The taxonomy of Loboscelidiinae in Vietnam is revised, with 16 new species being described: Loboscelidia bachmaensis sp. nov., L. barbata sp. nov., L. cilia sp. nov., L. convexa sp. nov., L. cucphuongensis sp. nov., L. cuneata sp. nov., L. do sp. nov., L. flavipes sp. nov., L. glabra sp. nov., L. komedai sp. nov., L. mediata sp. nov., L. parallela sp. nov., L. piriformis sp. nov., L. squamosa sp. nov., L. vang sp. nov. and L. vietnamensis sp. nov. In total, 24 species of Loboscelidia are recognized in the fauna of Vietnam. Keys to Indo-Chinese male and world female of Loboscelidia are provided. A brief observation of the foraging behavior of L. squamosa sp. nov. is also reported. Host-carriage and subsequent host egg burying are considered primary nesting behaviors of solitary wasps.
Over the last three decades, countries across the Andean region have moved toward legal recognition of indigenous justice systems. This turn toward legal pluralism, however, has been and continues to be heavily contested. The working paper explores a theoretical perspective that aims at analyzing and making sense of this contentious process by assessing the interplay between conflict and (mis)trust. Based on a review of the existing scholarship on legal pluralism and indigenous justice in the Andean region, with a particular focus on the cases of Bolivia and Ecuador, it is argued that manifest conflict over the contested recognition of indigenous justice can be considered as helpful and even necessary for the deconstruction of mistrust of indigenous justice. Still, such conflict can also help reproduce and even reinforce mistrust, depending on the ways in which conflict is dealt with politically and socially. The exploratory paper suggests four proposition that specify the complex and contingent relationship between conflict and (mis)trust in the contested negotiation of pluralist justice systems in the Andean region.
The interaction between Λ baryons and kaons/antikaons is a crucial ingredient for the strangeness S=0 and S=−2 sector of the meson−baryon interaction at low energies. In particular, the ΛK¯¯¯¯ might help in understanding the origin of states such as the Ξ(1620), whose nature and properties are still under debate. Experimental data on Λ−K and Λ−K¯¯¯¯ systems are scarce, leading to large uncertainties and tension between the available theoretical predictions constrained by such data. In this Letter we present the measurements of Λ−K+⊕Λ¯¯¯¯−K− and Λ−K−⊕Λ¯¯¯¯−K+ correlations obtained in the high-multiplicity triggered data sample in pp collisions at s√=13 TeV recorded by ALICE at the LHC. The correlation function for both pairs is modeled using the Lednicky−Lyuboshits analytical formula and the corresponding scattering parameters are extracted. The Λ−K−⊕Λ¯¯¯¯−K+ correlations show the presence of several structures at relative momenta k∗ above 200 MeV/c, compatible with the Ω baryon, the Ξ(1690), and Ξ(1820) resonances decaying into Λ−K− pairs. The low k∗ region in the Λ−K−⊕Λ¯¯¯¯−K+ also exhibits the presence of the Ξ(1620) state, expected to strongly couple to the measured pair. The presented data allow to access the ΛK+ and ΛK− strong interaction with an unprecedented precision and deliver the first experimental observation of the Ξ(1620) decaying into ΛK−.
In this work, inhomogeneous chiral phases are studied in a variety of Four-Fermion and Yukawa models in 2+1 dimensions at zero and non-zero temperature and chemical potentials. Employing the mean-field approximation, we do not find indications for an inhomogeneous phase in any of the studied models. We show that the homogeneous phases are stable against inhomogeneous perturbations. At zero temperature, full analytic results are presented.
In this work we study the 3+1-dimensional Nambu-Jona-Lasinio (NJL) model in the mean field-approximation. We carry out calculations using five different regularization schemes (two continuum and three lattice regularization schemes) with particular focus on inhomogeneous phases and condensates. The regularization schemes lead to drastically different inhomogeneous regions. We provide evidence that inhomogeneous condensates appear for all regularization schemes almost exclusively at values of the chemical potential and with wave numbers, which are of the order of or even larger than the corresponding regulators. This can be interpreted as indication that inhomogeneous phases in the 3+1-dimensional NJL model are rather artifacts of the regularization and not a consequence of the NJL Lagrangian and its symmetries.
Asymptotic giant branch (AGB) stars are responsible for the production of the main component of the solar s-process distribution. Despite enormous progress in the theoretical modeling of these objects over the last few decades, many uncertainties remain. The still-unknown mechanism leading to the production of 13C neutron source is one example. The nucleosynthetic signature of AGB stars can be examined in a number of stellar sources, from spectroscopic observations of intrinsic and extrinsic stars to the heavy-element isotopic composition of presolar grains found in meteorites. The wealth of available observational data allows for constraining the processes occurring in AGB interiors. In this view, we discuss recent results from new AGB models including the effects of mixing triggered by magnetic fields, and show comparisons of the related s-process nucleosynthesis with available observations.

The production of π±, K±, and (p¯¯¯)p is measured in pp collisions at s√=13 TeV in different topological regions. Particle transverse momentum (pT) spectra are measured in the ``toward'', ``transverse'', and ``away'' angular regions defined with respect to the direction of the leading particle in the event. While the toward and away regions contain the fragmentation products of the near-side and away-side jets, respectively, the transverse region is dominated by particles from the Underlying Event (UE). The relative transverse activity classifier, RT=NT/⟨NT⟩, is used to group events according to their UE activity, where NT is the measured charged-particle multiplicity per event in the transverse region and ⟨NT⟩ is the mean value over all the analysed events. The first measurements of identified particle pT spectra as a function of RT in the three topological regions are reported. The yield of high transverse momentum particles relative to the RT-integrated measurement decreases with increasing RT in both the toward and away regions, indicating that the softer UE dominates particle production as RT increases and validating that RT can be used to control the magnitude of the UE. Conversely, the spectral shapes in the transverse region harden significantly with increasing RT. This hardening follows a mass ordering, being more significant for heavier particles. The pT-differential particle ratios (p+p¯¯¯)/(π++π−) and (K++K−)/(π++π−) in the low UE limit (RT→0) approach expectations from Monte Carlo generators such as PYTHIA 8 with Monash 2013 tune and EPOS LHC, where the jet-fragmentation models have been tuned to reproduce e+e− results.
Graph data is an omnipresent way to represent information in machine learning. Especially, in neuroscience research, data from Diffusion-Tensor Imaging (DTI) and functional Magnetic Resonance Imaging (fMRI) is commonly represented as graphs. Exploiting the graph structure of these modalities using graph-specific machine learning applications is currently hampered by the lack of easy-to-use software. PHOTONAI Graph aims to close the gap between domain experts of machine learning, graph experts and neuroscientists. Leveraging the rapid machine learning model development features of the Python machine learning API PHOTONAI, PHOTONAI Graph enables the design, optimization, and evaluation of reliable graph machine learning models for practitioners. As such, it provides easy access to custom graph machine learning pipelines including, hyperparameter optimization and algorithm evaluation ensuring reproducibility and valid performance estimates. Integrating established algorithms such as graph neural networks, graph embeddings and graph kernels, it allows researchers without significant coding experience to build and optimize complex graph machine learning models within a few lines of code. We showcase the versatility of this toolbox by building pipelines for both resting–state fMRI and DTI data in the hope that it will increase the adoption of graph-specific machine learning algorithms in neuroscience research.
Quantum-correlated DD¯ pairs collected by the BESIII experiment at the ψ(3770) resonance, corresponding to an integrated luminosity of 2.93 fb−1, are used to study the D0→K0Sπ+π−π0 decay mode. The CP-even fraction of D0→K0Sπ+π−π0 decays is determined to be 0.235±0.010±0.002, where the first uncertainty is statistical and the second is systematic.
How does group identity affect belief formation? To address this question, we conduct a series of online experiments with a representative sample of individuals in the US. Using the setting of the 2020 US presidential election, we find evidence of intergroup preference across three distinct components of the belief formation cycle: a biased prior belief, avoid-ance of outgroup information sources, and a belief-updating process that places greater (less) weight on prior (new) information. We further find that an intervention reducing the salience of information sources decreases outgroup information avoidance by 50%. In a social learn-ing context in wave 2, we find participants place 33% more weight on ingroup than outgroup guesses. Through two waves of interventions, we identify source utility as the mechanism driving group effects in belief formation. Our analyses indicate that our observed effects are driven by groupy participants who exhibit stable and consistent intergroup preferences in both allocation decisions and belief formation across all three waves. These results suggest that policymakers could reduce the salience of group and partisan identity associated with a policy to decrease outgroup information avoidance and increase policy uptake.
Objectives In this early retrospective cohort study, a total of 26 patients with SARS-CoV-2 were treated with bamlanivimab or casirivimab/imdevimab, and the reduction of the viral load associated with the developed clinical symptoms was analyzed.
Methods: Patients in the intervention groups received bamlanivimab or casirivimab/imdevimab. Patients without treatment served as control. Outcomes were assessed by clinical symptoms and change in log viral load from baseline based on the cycle threshold over a period of 18 days.
Results: Median log viral load decline was higher in both intervention groups after 3 and 6 days compared to control. However, at later time points, the decline of the viral load was more distinct in the control group. Mild symptoms of COVID-19 were observed in 6.3% of the intervention groups and in no patient of the control. No patients treated with bamlanivimab, 18.8% treated with casirivimab/imdevimab, and 14.2% in the control group developed moderate symptoms. Severe symptoms were recorded only in the control group (14.2%), including one related death.
Conclusion: Treatment with monoclonal SARS-CoV-2 antibodies seems to accelerate decline of virus loads, especially in the first 6 days after administration, compared to control. This may be associated with a reduced likeliness of a severe course of COVID-19.
In this paper, I will address the issue of translation as a critique of autochthony that emerges in the context of Fritz Mauthner's linguistic scepticism. Translation, for Mauthner, becomes a privileged prism through which to consider identity and belonging, as well as a way of understanding uprootedness, since language is a continuous product of borrowing, bastardization, stratification, and contingency. According to Mauthner, languages are not possession, but borrowing; not purity, but contagion; not an abstract crystallization, but transit. Therefore, love of the mother tongue - the only way to conceive patriotism - is not a physical connection with the land, roots, or nation, but a refuge, an always precarious 'Heimat' (home).
The more difficult it is to access a research field, the more substantial the need to develop creative methodological models. Investigating the psychosocial impact of migration-related carceral spaces constitutes one such research field. To shed light on these spaces and counter the challenges of opacity, harm, and power asymmetries, we propose a psycho-geographical counter-mapping as a mixed methods approach based on grounded theory (MM-GT). Applying it to immigration detention and refugee confinement sites in Mexico and Samos, our analysis proposes a fully integrated and sequential design that can be adapted to a range of carceral spaces and subjectivation in transdisciplinary research.

The purpose of this study was to examine if prosodic patterns in oral reading derived from Recurrence Quantification Analysis (RQA) could distinguish between struggling and skilled German readers in Grades 2 (n = 67) and 4 (n = 69). Furthermore, we investigated whether models estimated with RQA measures outperformed models estimated with prosodic features derived from prosodic transcription. According to the findings, struggling second graders appear to have a slower reading rate, longer intervals between pauses, and more repetitions of recurrent amplitudes and pauses, whereas struggling fourth graders appear to have less stable pause patterns over time, more pitch repetitions, more similar amplitude patterns over time, and more repetitions of pauses. Additionally, the models with prosodic patterns outperformed models with prosodic features. These findings suggest that the RQA approach provides additional information about prosody that complements an established approach.

In natural environments, background noise can degrade the integrity of acoustic signals, posing a problem for animals that rely on their vocalizations for communication and navigation. A simple behavioral strategy to combat acoustic interference would be to restrict call emissions to periods of low-amplitude or no noise. Using audio playback and computational tools for the automated detection of over 2.5 million vocalizations from groups of freely vocalizing bats, we show that bats (Carollia perspicillata) can dynamically adapt the timing of their calls to avoid acoustic jamming in both predictably and unpredictably patterned noise. This study demonstrates that bats spontaneously seek out temporal windows of opportunity for vocalizing in acoustically crowded environments, providing a mechanism for efficient echolocation and communication in cluttered acoustic landscapes.
One Sentence Summary: Bats avoid acoustic interference by rapidly adjusting the timing of vocalizations to the temporal pattern of varying noise.
The ALICE Collaboration reports measurements of the semi-inclusive distribution of charged-particle jets recoiling from a high transverse momentum (high pT) charged hadron, in pp and central Pb−Pb collisions at center of mass energy per nucleon−nucleon collision sNN−−−√=5.02 TeV. The large uncorrelated background in central Pb−Pb collisions is corrected using a data-driven statistical approach, which enables precise measurement of recoil jet distributions over a broad range in pT,chjet and jet resolution parameter R. Recoil jet yields are reported for R=0.2, 0.4, and 0.5 in the range 7<pT,chjet<140 GeV/c and π/2<Δφ<π, where Δφ is the azimuthal angular separation between hadron trigger and recoil jet. The low pT,chjet reach of the measurement explores unique phase space for studying jet quenching, the interaction of jets with the quark-gluonnplasma generated in high-energy nuclear collisions. Comparison of pT,chjet distributions from pp and central Pb−Pb collisions probes medium-induced jet energy loss and intra-jet broadening, while comparison of their acoplanarity distributions explores in-medium jet scattering and medium response. The measurements are compared to theoretical calculations incorporating jet quenching.
Binary neutron star mergers represent unique observational phenomena because all four fundamental interactions play an important role at various stages of their evolution by leaving imprints in astronomical observables. This makes their accurate numerical modeling a challenging multiphysics problem that promises to increase our understanding of the high-energy astrophysics at play, thereby providing constraints for the underlying fundamental theories such as the gravitational interaction or the strong interaction of dense matter. For example, the first and so far only multi-messenger observation of the binary neutron star merger GW170817 resulted in numerous bounds on the parameters of isolated non-rotating neutron stars, e.g., their maximum mass or their distribution in radii, which can be directly used to constrain the equation of state of cold nuclear matter. While many of these results stem from the observation of the inspiral gravitational-wave signal, the postmerger phase of binary neutron star mergers encodes even more details about the extreme physics of hot and dense neutron star matter. In this Thesis we focus on the exploration of dissipative and shearing effects in binary neutron star mergers in order to identify novel approaches to constrain hot and dense neutron star matter.
The first effect is the well-motivated dissipation of energy due to the bulk viscosity which arises from violations of weak chemical equilibrium. We start by exploring the impact of bulk viscosity on black-hole accretion. This simplified problem gives us the opportunity to develop a test case for future codes taking into account the effects of dissipation in a fully general-relativistic setup and build intuition in the physics of relativistic dissipation. Next, we move on to isolated neutron stars and binary neutron star mergers by developing a robust implementation of bulk-viscous dissipation for numerical relativity simulations. We test our implementation by calculating the damping of eigenmodes of isolated neutron stars and the violent migration scenario. Finally, we present the first results on the impact of bulk viscosity on binary neutron star mergers. We identify a number of ways how bulk viscosity impacts the postmerger phase, out of which the suppression of gravitational-wave emission and dynamical mass ejection are the most notable ones.
In the last part of this Thesis we investigate how the shearing dynamics at the beginning of the merger affects the amplification of different initial magnetic-field topologies. We explore the hypothesis that magnetic fields which are located only in a small region near the stellar surface prior to merger lead to a weaker magnetic-field amplification. We show first evidence which confirms this hypothesis and discuss possible implications for constraining the physics of superconduction in cold neutron stars.
Hintergrund: Epilepsie bezeichnet eine Erkrankung, welche durch eine anhaltende Prädisposition für Symptome, die im Zusammenhang mit einer ungewöhnlich starken oder synchronisierten elektrischen Aktivität des Gehirns auftreten (=epileptischer Anfall) charakterisiert ist. Mutationen in dem Gen DEPDC5 (Dishevelled, Egl-10 and Pleckstrin (DEP) domain-containing protein 5) sind mit fokalen Epilepsien assoziiert und führen in Tiermodellen und humanen Modellen zu einer Überaktivierung des mTOR-Signalweges. Auf neuromorphologischer Ebene zeigt sich die mTOR-Überaktivierung durch eine Vergrößerung des Zelldurchmessers und einer zunehmenden Verästelung der Neuriten. Ziel dieser Studie war es, die morphologischen Auswirkungen der DEPDC5-assoziierten mTOR-Überaktivierung in der SH-SY5Y-Neuroblastomzelllinie zu untersuchen. Dadurch soll eine Einschätzung getroffen werden, ob das im Zusammenhang mit neurodegenerativen Erkrankungen bereits gut etablierte SH-SY5Y-Zellmodell auch bei der Untersuchung epilepsieassoziierter Pathomechanismen zum Einsatz kommen kann.
Methoden: Unter Einsatz der CRISPR/Cas9-Methode wurden Knockout(KO)-Mutationen in Exon 2 und Exon 3 des DEPDC5-Gens erzeugt und diese mittels Sanger-Sequenzierung bestätigt. Danach wurden die Knockouts auf RNA- und Proteinebene, durch Real-time-RT-PCR und Western Blot validiert. Die bestätigten homozygoten DEPDC5-KO-Zelllinen wurden anschließend mittels Western Blot auf eine mTOR-Überaktivierung untersucht. Zuletzt erfolgte die neuromorphologische Validierung des DEPDC5-KO. Die Zellgröße proliferierender SH-SY5Y wurden mittels Durchflusszytometrie (FACS) untersucht. Zudem bestimmten wir die neuronale Architektur differenzierter SH-SY5Y unter Einsatz der Sholl-Analyse.
Ergebnisse: Es konnten vier unabhängige DEPDC5-KO-SH-SY5Y-Zelllinien mit homozygoten Indel-Mutationen und vorzeitigem Stoppcodon in Exon 3 generiert werden. Die erwartete Reduktion an DEPDC5-mRNA konnte mittels Real-time 10 RT-PCR nicht festgestellt werden. Die Abwesenheit des Proteins konnte durch Western Blot aber gezeigt werden. Funktionell konnte für alle Zelllinien eine mTOR-Überaktivierung mittels Western Blot nachgewiesen werden. Dabei konnte phosphoryliertes AKT (AKT serine/threonine kinase 1) als stabilster Marker etabliert werden. Auf neuromorphologischer Ebene ließ sich ein Trend in Richtung vergrößertem Zelldurchmesser bei verlängertem Auswachsen der Neuriten feststellen, wobei sich für das Modell Unterschiede zwischen den einzelnen Klonen ergaben.
Diskussion: In dieser Studie gelang es erstmals, den Zusammenhang zwischen DEPDC5-KO und einer mTOR-Überaktivierung in der onkogenen SH-SY5Y-Zelllinie zu replizieren. Das verlängerte Auswachsen der Neuriten, bei jedoch gleichbleibender Anzahl peripherer Verästelungen, stellt dabei einen neuen Befund dar und könnte durch die frühe neuronale Entwicklungsstufe des SH-SY5Y-Zellmodells erklärt werden. Auf Grundlage der Ergebnisse dieser Arbeit lässt sich sagen, dass das robuste und kostengünstige SH-SY5Y-Zellmodell insbesondere für high-throughput Methoden und Screeningassays ein geeignetes Modell ist. Durch die Kombination mit reiferen Zellmodellen, wie beispielsweise iPSCs (induced pluripotent stem cells), könnte der Phänotyp eines DEPDC5-KO und anderer mTOR-assoziierter Epilepsien, möglichst umfassend in-vitro dargestellt werden.

Mit der Figur Belacane präsentiert Wolfram von Eschenbach im "Parzival" seinem Publikum eine Frau, die den höfischen Schönheitsvorstellungen widerspricht. Immer wieder wird die Schwarze Hautfarbe Belacanes und ihres Volkes explizit im Kontrast zum dagegen unmarkierten weißen Protagonisten betont und abgewertet. Erst die Information über Belacanes Jungfräulichkeit macht sie für den christlichen Ritter Gahmuret attraktiv, der ihre Tugendhaftigkeit als Zeichen ihrer innerlichen Taufe sieht. Dass Belacane unter all den Menschen, die "höllenfarben" (51,24) sind, eine Ausnahmestellung zugesprochen wird, offenbart die Abwertung aller anderen BIPoC und macht sie für Gahmuret zum Objekt der Begierde. Er befreit die Jungfrau in Nöten erst von ihren Belagerern, dann von ihrer Keuschheit und wird so zum König über ihr Land, bis er die schwangere Belacane plötzlich heimlich verlässt. Der Brief, den er ihr hinterlässt, dokumentiert die Mehrfachabwertung Belacanes: aufgrund ihres Frau-Seins wird sie zum Medium der Stammeserhaltung, aufgrund ihres zukünftigen Mutter-Seins zum Kommunikationsmittel mit dem noch ungeborenen Sohn und aufgrund ihres heidnischen Glaubens zur Legitimation für die Trennung. Hannah Mieger analysiert anhand der Belacane-Episode, wie die drei Differenzkategorien Race, Religion und Gender miteinander interagieren und zeigt dadurch, dass prämoderne Texte intersektional lesbar sind.
Recent empirical evidence shows that most international prices are sticky in dollars. This paper studies the policy implications of this fact in the context of an open economy model, allowing for an arbitrary structure of asset markets, general preferences and technologies, time- or state-dependent price setting, and a rich set of shocks. We show that although monetary policy is less efficient and cannot implement the flexible-price allocation, inflation targeting remains robustly optimal in non-U.S. economies. The implementation of this non-cooperative policy results in a "global monetary cycle" with other countries importing the monetary stance of the U.S. The capital controls cannot unilaterally improve the allocation and are useful only when coordinated across countries. Thanks to the dominance of the dollar, the U.S. can extract rents in international goods and asset markets and enjoy a higher welfare than other economies. Although international cooperation benefits other countries by improving global demand for dollar-invoiced goods, it is not in the self-interest of the U.S. and may be hard to sustain.

Xylose, an abundant sugar fraction of lignocellulosic biomass, is a five-carbon skeleton molecule. Since decades, utilization of this sugar has gained much attention and has been in particular focus as a substrate for production of biofuels like ethanol by microbial hosts, including Saccharomyces cerevisiae. In this yeast, xylose is naturally not used as a carbon source, but its utilization could be achieved by metabolic engineering either via the oxidoreductive route or through the isomerase pathway. Both pathways share xylulose as a common intermediate that must be phosphorylated before entering the endogenous metabolism via the non-oxidative pentose phosphate pathway (noxPPP). Besides this, in some bacteria a non-phosphorylating oxidative pathway for xylose degradation exists, known as Weimberg pathway, where a molecule of xylose is converted by a series of enzymes - xylose dehydrogenase (XylB), xylonate dehydratase (XylD), 3-keto-2-deoxy-xylonate dehydratase (XylX) and α-ketoglutarate semialdehyde dehydrogenase (KsaD) - to form α-ketoglutarate (AKG). Besides having several useful properties as a product, AKG could also be used for cell growth as an intermediate of the tricarboxylic acid (TCA) cycle. One target of the present study is to establish a functional Weimberg pathway in S. cerevisiae. Previous studies have shown that this task is not trivial, for instance due to the toxicity of xylonate (the first metabolite of the pathway) and the involvement of an iron-sulfur cluster dependent enzyme, the D-xylonate dehydratase. The assembly of iron-sulfur clusters on a heterologous protein in yeast is known to be challenging.
To establish the Weimberg pathway in yeast, the genes xylB, xylD, and xylX were obtained from Caulobacter cresentus and ksaD was from Corynebacterium glutamicum. In a variant, the dehydratase xylD was replaced with orf41 from Arthrobacter nicotinovorans, which is believed to be independent of iron-sulfur clusters. Growth of yeast cells on xylose as a sole carbon source was expected as an indicator of a functional Weimberg pathway. However, the heterologous expression of the codon optimized genes was not sufficient to reach this goal. Due to the complexity of the interactions of the heterologous pathway with the endogenous cellular processes, it was assumed that potential limitations could be overcome by adaptive laboratory evolution, using xylose as a sole source of carbon. Increasing selection pressure was applied on a strain with Weimberg pathway genes integrated into the genome over several generations. As a variant of the evolutionary engineering approach, mutator strains were generated. For this, RAD27 and MSH2 genes were deleted, which are involved in nucleotide excision and mismatch repair mechanisms, respectively. Some of the resulting strains PRY24, PRY25, PRY27 and PRY28 were able grow in xylose as a sole carbon source after evolutionary engineering. As a control, a non-mutator strain PRY19 was also included. Strikingly, only the mutator strains were able to consume xylose as a sole carbon source, which shows the feasibility of the approach.
In addition to the mutator strain strategy, a further approach employed in the present study was the simultaneous expression of the Weimberg pathway in the cytosol and mitochondria. This was based on the reasoning that the iron-sulfur cluster biogenesis on XylD may be improved in the organelle and that the AKG is an intermediate of the TCA cycle. In the strain AHY02, all enzymes of the pathway were tagged with mitochondrial targeting signals in addition to a full cytosolically localized pathway. The localization of the mitochondrial variants was confirmed by fluorescence microscopy. Together with AHY02, CEN.PK2-1C wild type strain was also included as a control for evolution. When a selection pressure on xylose was applied, both strains - AHY02 and CEN.PK2-1C - were able to grow in the course of evolution. Deletion of the xylulokinase (XKS1) gene was found to be detrimental for both evolved strains in xylose-containing media. This suggests that the evolution of the endogenous oxidoreductive and noxPPP genes is responsible for growth of the evolved cells. For the evolved strain AHY02, it could also be possible that the Weimberg pathway genes supported to growth in addition to the oxidoreductive route. To elucidate the underlying molecular mechanisms, genome sequencing and reverse engineering approaches would be necessary in future.
In addition to screening for growth on xylose as a sole carbon source, a less stringent screening system was created to examine even a minor flux of xylose towards AKG. For this, all genes necessary for conversion of isocitrate to AKG where deleted, yielding a glutamate auxotrophic strain. Trauma ist in Deutschland und weltweit eine der häufigsten Todesursache bei Personen unter 55 Jahren. Eine traumatische Verletzung von Gewebe führt zur Freisetzung von sogenannten damage-associated molecular patterns (DAMPs), die eine Entzündungskaskade auslösen, welche die Organfunktion negativ beeinträchtigt. Dies führt bei anhaltender Entzündung zunächst zu Organdysfunktion, was im weiteren Verlauf zu Organversagen führt und im Spätstadium im multiplen Organversagen (MOF) enden kann. In den meisten Fällen ist die inflammatorische Antwort des Immunsystems auf das Trauma adäquat und entspricht einem gut koordinierten Netzwerk von Immunzellen, Zytokinen und Chemokinen, welches zur Wiederherstellung des geschädigten Gewebes führt. Wenn dieses Netzwerk jedoch nicht im Gleichgewicht ist, kann die Entzündungsreaktion durch eine sogenannte feed-forward-loop von Inflammation und Gewebeschäden verstärkt werden. Wenn dieser Prozess systemisch wird, spricht man vom systemic inflammatory response syndrome (SIRS). Auf der anderen Seite gibt es ein Gegenstück zu SIRS, nämlich das sogenannte compensatory anti-inflammatory response syndrome (CARS). Auch ein Überschießen von CARS kann den Verlauf der posttraumatischen Inflammation negativ beeinträchtigen. Sowohl eine Verschiebung in Richtung von SIRS als auch in Richtung von CARS kann zu Organfunktionsstörungen, nosokomialen Infektionen und letztendlich zum Tod führen. Daher ist eine ausgeglichene, frühe posttraumatische Immunantwort für ein gutes Outcome von entscheidender Bedeutung.
Monozyten nehmen eine kritische Stellung sowohl in der primären Immunantwort nach Trauma als auch nach Infektion ein. Durch die Oberflächenexpression von diversen sogenannten pattern-recocnition receptors (PRRs), insbesondere Toll-Like-Rezeptoren (TLRs), können Monozyten Pathogene und sogenannte pathogen-associated molecular patterns (PAMPs) erkennen und neutralisieren. Darüber hinaus können Monozyten PAMPs über major histocompatibility complex class II -Moleküle (MHC-II) dem adaptiven Immunsystem präsentieren und somit als zelluläre Verbindung zwischen dem angeborenen und dem adaptiven Immunsystem fungieren. TLR2, eine Untergruppe der TLRs, ist einer der Hauptrezeptoren für PAMPs von grampositiven Bakterien wie S. aureus, dem Hauptkeim für posttraumatische Wundinfektionen und nosokomiale Infektionen.
Es gibt immer noch keinen Konsens über den Einfluss von Trauma auf die Funktion von Monozyten und deren Expressionsprofil von PRRs und MHC-II-Molekülen nach einem Trauma. Verschiedene Studien berichten von einer beeinträchtigten TLR2-Expression bei Monozyten nach Trauma, während sich in anderen Studien eine konstante oder sogar erhöhte TLR2-Expression bei Monozyten nach Trauma gezeigt hat. Die Daten sind auch in Bezug auf die posttraumatische Phagozytoseleistung von Monozyten unbeständig. Während in einigen Studien im frühen posttraumatischen Verlauf im Vergleich zu gesunden Probanden von einer erhöhten Anzahl zirkulierender Monozyten mit erhaltener Phagozytoseleistung berichtet wird, zeigten andere Studien eine verminderte Phagozytoseleistung von Monozyten nach Trauma. Darüber hinaus gibt es widersprüchliche Ergebnisse bezüglich der Fähigkeit von Monozyten, proinflammatorische Zytokine wie Interleukin (IL)-1β oder Tumor necrosis factor (TNF)-α freizusetzen.
Zusammengefasst ist die derzeitige Studienlage widersprüchlich. Zusätzlich wurde es in bisherigen Studien verpasst, die Kombination verschiedener Aspekte wie die Funktionalität der Monozyten in Bezug auf Phänotypisierung oder funktioneller Assays und deren Beobachtung über einen längeren Zeitraum miteinzubeziehen.
Die unterschiedlichen Ergebnisse der bisherigen Studien könnte durch die Natur des Polytraumas bedingt sein, beispielsweise durch verschiedene Verletzungsmuster und Verletzungsschwere, die unterschiedliche Erstversorgung am Unfallort, die Zeit bis zur Ankunft in der Notaufnahme oder die unterschiedliche primäre und definitive Versorgung der Verletzungen. All das kann zu einer unterschiedlichen Antwort des Immunsystems und somit zu unterschiedlichen Outcomes führen.
Daher war es Ziel unserer Studie, die oben genannten Variablen zu eliminieren, und eine kontrollierte Polytraumastudie am Schweinemodell durchzuführen, um die Funktionalität von Schweinemonozyten über einen Zeitraum von 72 Stunden nach Trauma zu untersuchen.
Zusätzlich und im Gegensatz zu früheren Studien haben wir die Phänotypisierung von Monozyten (TLR2 und MHC-II [SLA-DR]) mit einem funktionellen Phagozytose-Assay kombiniert und deren direkte Assoziation in einem unabhängigen Assay analysiert.
Peripheres Blut wurde vor (-1h) und direkt nach der Induktion des Polytraumas (PT) (0h) entnommen, sowie 3,5h, 5,5h, 24h und 72h später. Die Expression von H(S)LA-DR und TLR2 auf Schweine-Monozyten wurde untersucht. Außerdem wurde die Phagozytierungsaktivität von Schweinemonozyten gemessen.
Darüber hinaus wurden aus mechanistischen Gründen Blutproben von 10 gesunden Schweinen zunächst einem TLR2-neutralisierenden Antikörper und anschließend S. aureus-Partikeln ausgesetzt, bevor die Phagozytoseleistung der Monozyten untersucht wurde.
Die Anzahl der CD14 + -Monozyten aller zirkulierenden Leukozyten blieb während des Beobachtungszeitraums konstant, während der Prozentsatz der CD14 + H(S)LA-DR + -Monozyten direkt, 3,5h und 5,5h nach dem Trauma signifikant abnahm. Der Prozentsatz von TLR2 + exprimierenden Zellen aus allen Monozyten verringerte sich direkt, 3,5h und 5,5h nach dem Trauma signifikant. Der Prozentsatz der phagozytierenden Monozyten nahm direkt nach Trauma ab und blieb in den ersten 3,5 Stunden nach dem Trauma niedriger, stieg jedoch nach 24 Stunden an. Die Antagonisierung von TLR2 verringerte signifikant die Phagozytoseleistung der Monozyten.
Sowohl der verringerte Prozentsatz der aktivierten als auch der TLR2-exprimierenden Monozyten blieb bestehen, solange die verringerte Phagozytoseleistung beobachtet wurde.
Darüber hinaus führte auch die Neutralisation von TLR2 zu einer verminderten Phagozytoseleistung. Daher nehmen wir an, dass eine verringerte TLR2-Expression für die verringerte Phagozytoseleistung verantwortlich ist.
In QCD at large enough isospin chemical potential Bose-Einstein Condensation (BEC) takes place, separated from the normal phase by a phase transition. From previous studies the location of the BEC line at the physical point is known. In the chiral limit the condensation happens already at infinitesimally small isospin chemical potential for zero temperature according to chiral perturbation theory. The thermal chiral transition at zero density might then be affected, depending on the shape of the BEC boundary, by its proximity. As a first step towards the chiral limit, we perform simulations of 2+1 flavors QCD at half the physical quark masses. The position of the BEC transition is then extracted and compared with the results at physical masses.
Scelolabes, historically is a monotypic genus of Ocydromiinae (Hybotidae) that for a long time was not studied due to the absence of the type of the type species (Scelolabes bivittatus Philippi), which is probably lost or destroyed. In this context, we propose an updated diagnosis and redescription of the genus, based on specimens identified by experts and in agreement with the original description of Philippi (1865), as well as illustrating male and female terminalia for the first time. This serves as an aid to discuss and corroborate the status of Scelolabes in the Neotropical region as a genus distinct from Hoplopeza. In addition, two new species are described, and an identification key and a distribution map are provided to all the Neotropical species of the genus.
Approximately 80 % of persistent wound infections are affected by the presence of bacterial biofilms, resulting in a severe clinical challenge associated with prolonged healing periods, increased morbidity, and high healthcare costs. Unfortunately, in vitro models for wound infection research almost exclusively focus on early infection stages with planktonic bacteria. In this study, we present a new approach to emulate biofilm-infected human wounds by three-dimensional human in vitro systems. For this purpose, a matured biofilm consisting of the clinical key wound pathogen Pseudomonas aeruginosa was pre-cultivated on electrospun scaffolds allowing for non-destructive transfer of the matured biofilm to human in vitro wound models. We infected tissue-engineered human in vitro skin models as well as ex vivo human skin explants with the biofilm and analyzed structural tissue characteristics, biofilm growth behavior, and biofilm-tissue interactions. The structural development of biofilms in close proximity to the tissue, resulting in high bacterial burden and in vivo-like morphology, confirmed a manifest wound infection on all tested wound models, validating their applicability for general investigations of biofilm growth and structure. The extent of bacterial colonization of the wound bed, as well as the subsequent changes in molecular composition of skin tissue, were inherently linked to the characteristics of the underlying wound models including their viability and origin. Notably, the immune response observed in viable ex vivo and in vitro models was consistent with previous in vivo reports. While ex vivo models offered greater complexity and closer similarity to the in vivo conditions, in vitro models consistently demonstrated higher reproducibility. As a consequence, when focusing on direct biofilm-skin interactions, the viability of the wound models as well as their advantages and limitations should be aligned to the particular research question of future studies. Altogether, the novel model allows for a systematic investigation of host-pathogen interactions of bacterial biofilms and human wound tissue, also paving the way for development and predictive testing of novel therapeutics to combat biofilm-infected wounds.

Die Welt, die Annette Werner untersucht, erscheint uns fremd, fast schon absurd: Verschiedene Zahlen haben hier die gleiche Größe, und Kreise besitzen unendlich viele Mittelpunkte. Die Mathematikprofessorin forscht auf dem Gebiet der sogenannten p-adischen Geometrie – einem Bereich der modernen Algebra, der in den letzten Jahrzehnten einen stürmischen Fortschritt erlebt hat.
Gene therapy has garnered increasing interest over recent decades. Several therapies employing gene transfer mechanisms have been developed, and, of these, adeno-associated virus (AAV) vectors have demonstrated viability for use with in vivo gene therapy. Several AAV-based therapeutics have received regulatory approval in the last few years including those for retinal disease, spinal muscular atrophy or aromatic L-amino acid decarboxylase deficiency. Lately, with the introduction of novel liver-directed AAV vector-based therapeutics for the treatment of haemophilia A and B, gene therapy has attracted significant attention in the hepatology community, with the liver increasingly recognised as a target for gene therapy. However, the introduction of foreign DNA into hepatocytes is associated with a risk of hepatic reactions, with raised ALT (alanine aminotransferase) and AST (aspartate aminotransferase) being – so far – the most commonly reported side effects. The complete mechanisms underlying the ALT flairs remain to be determined and the long-term risks associated with these new treatments is not yet known. The liver community is increasingly being asked to support liver-directed gene therapy to mitigate potential liver associated harm. In this review, we focus on AAV vector-based gene therapy, shedding light on this promising technique and its remarkable success in haemophilia, with a special focus on hepatic complications and their management in daily clinical practice.
Despite the recent popularity of predictive processing models of brain function, the term prediction is often instantiated very differently across studies. These differences in definition can substantially change the type of cognitive or neural operation hypothesised and thus have critical implications for the corresponding behavioural and neural correlates during visual perception. Here, we propose a five-dimensional scheme to characterise different parameters of prediction. Namely, flow of information, mnemonic origin, specificity, complexity, and temporal precision. We describe these dimensions and provide examples of their application to previous work. Such a characterisation not only facilitates the integration of findings across studies, but also helps stimulate new research questions.
The lianescent genus Tinospora Miers in Singapore is revised. Four species are recognised, two of which are newly described. Tinospora krispura I.M.Turner sp. nov. is known from Christmas Island and Singapore. Tinospora singapura I.M.Turner sp. nov. is known only from Singapore. Two new lectotypifications are included, one at the second step.

This article reads Albert Cohen's "Le Livre de ma mère", which mourns the death of his mother, as a poetics of love and loss. It is a poetics of otherness that disavows the claim to expression and selfhood. The mother, being the paradigmatic figure of otherness, is a figure for literature, a form of language that is characterized by saying things differently. Literature itself is a motherly space insofar as it others the language of the self. This argument is developed along close readings of both the French original and the English translation of Cohen's work, following three thematic axes: first, the peculiar kinship of love and death; second, the mother as the other; third, literature as filio-logy: a logic of filiation that does not leave the self unchanged.
Der erste Teil der vorliegenden Arbeit beinhaltet die funktionelle Analyse von fünf Oberflächenproteinen von B. recurrentis die die Fähigkeit besitzen, die Aktivierung von humanen Komplement zu inhibieren und Borrelien vor Bakteriolyse zu schützen. Im zweiten Teil der Arbeit wurden zwei immunologische Testverfahren mit hoher Sensitivität sowie Spezifität entwickelt und mit zahlreichen Patientenseren evaluiert. Die entwickelten Tests könnten in Zukunft als zuverlässige Instrumente für eine gesicherte Diagnose von LRF eingesetzt werden.
Eine Sequenzanalyse führte zur Identifizierung eines neuen Proteinclusters, welches die fünf untersuchten Komplement-inhibierenden Proteine als „Cluster of Complement-targeting and Host-interacting Proteins“ oder „Chi-Gencluster“, zusammenfasst. Diese Oberflächenproteine wurden als ChiA, ChiB, ChiC, ChiD und ChiE bezeichnet. Weiterführende Sequenzanalysen ergaben, dass das Chi-Gencluster extrem hoch konserviert ist und sowohl in den ersten B. recurrentis-Isolaten aus den 1990er Jahren als auch in B. recurrentis-Stämmen nachgewiesen werden konnte, die 2015 aus Patienten isoliert wurden.
Durch funktionelle Analysen konnte gezeigt werden, dass alle fünf Chi-Proteine in der Lage sind den alternativen und terminalen Komplementweg zu inhibieren. Ebenfalls konnte für die Proteine ChiB, ChiD sowie ChiE nachgewiesen werden, dass die Interaktion mit der Komplementkomponente C5 dosisabhängig verläuft.
Die strukturelle Aufklärung des Proteins ChiB ermöglichte es Aminosäuren zu identifizieren, von denen angenommen wurde, dass sie für die Interaktion mit Komplement eine Rolle spielen könnten. Durch in vitro Mutagenese konnten insgesamt fünf verschiedene Varianten von ChiB generiert werden, die jedoch keine Veränderungen in ihrem Komplement-inhibierenden Potential gegenüber dem unveränderten ChiB-Protein aufwiesen. Weder in der Inhibition des alternativen oder des terminalen Komplementweges, noch in der Interaktion mit den untersuchten Komplementkomponenten C3b, C5 und C9.
Weiter konnte gezeigt werden, dass die lytische Aktivität von Humanserum durch Vorinkubation mit ChiB, ChiC, ChiD und ChiE drastisch reduziert werden konnte, sodass Serum-sensible Borrelienzellen in Gegenwart von Komplement überlebten. „Gain-of-function“ B. garinii-Transformanten, welche mit dem entsprechendem Chi-kodierenden Gen transformiert wurden, bestätigten die mit den gereinigten Proteinen erhobenen Ergebnisse.So konnte nachgewiesen werden, dass ChiB-, ChiC- oder ChiD-produzierende „Gain-of-function“ B. garinii Transformanten, nicht jedoch ChiE- produzierende Zellen, in der Lage waren einen Serum-resistenten Phänotypen auszubilden. Für Transformanten, die zwei-, drei- oder vier Chi-Proteine in verschiedenen Kombinationen gleichzeitig produzierten, konnte allerdings die Fähigkeit in Gegenwart von Humanserum zu überleben nicht bestätigt werden.
Molekulare Analysen mit verschiedenen RF-Borrelienstämmen führten zum Nachweis, dass die fünf Chi-kodierenden Gene bei allen Isolaten vorhanden sind und unter in vitro Bedingungen exprimiert werden. Im Gegensatz zu B. recurrentis PAbJ, ließ sich das HcpA kodierende Gen in B. duttonii LAI nicht nachweisen, jedoch alle dem Chi-Cluster zugehörigen Gene. Bei B. duttoni V fehlte das gesamte Chi-Cluster sowie die für CihC- und HcpA-kodierenden Gene. Durch eine Western Blot-Analyse konnte mit spezifischen Antikörpern bestätigt werden, dass die Proteine CihC, HcpA und ChiB in B. recurrentis A17 unter in vitro Bedingungen produziert wurden.
Im zweiten Teil der vorliegenden Arbeit wurden durch die Analyse der IgM- und IgG-Immunreaktivitäten der LRF-Patientenseren zwei Proteine identifiziert, CihC und GlpQ, die als potenzielle Antigene für die Serodiagnostik des LRF evaluiert wurden. Eine initiale Evaluierung des IgM Lineblot-Immmunoassays zeigte jedoch nur eine geringe Sensitivität für die beiden Antigene, während der IgG Lineblot-Immunoassay eine sehr hohe Sensitivität aufwies. Der ELISA hingegen zeigte bei einer Kombination beider Antigene sehr gute Sensitivitäten und Spezifitäten. Um die starke Hintergrundfärbung bei den Lineblot-Immunoassays, welche eine korrekte Bewertung der Reaktivitäten gegenüber CihC erheblich erschwerten, zu minimieren, wurde ein „Epitop-Mapping“ durchgeführt, um immunogene Regionen innerhalb des CihC-Proteins zu lokalisieren. Eine zweite Evaluierung mit dem immunreaktiven N-terminalen CihC-Fragment CihC-N führte zu einer deutlichen Verbesserung der IgG Lineblot-Immunoassays mit einer Sensitivität von 100 % und einer starken Reduktion der Hintergrundfärbung. Zusätzlich konnte die Sensitivität der IgM-ELISA deutlich verbessert werden. Die Verwendung von CihC-N führte beim IgG-ELISA zur Herabsetzung des Cut-off-Wertes und zu einer besseren Unterscheidung zwischen den positiven LRF-Seren und den verwendeten Kontrollseren. Im Rahmen dieser Arbeit konnten somit zwei serologische in vitro Diagnostika entwickelt werden, die als zuverlässige Point-of-Care-Diagnostik in klinischen Studien eingesetzt werden könnten. Zur Steigerung der Sensitivität des IgM-Lineblot-Immunoassays sollten allerdings weiterführende Untersuchungen mit weiteren immunreaktiven Antigenen, wie z.B. den Vmp-Proteinen von B. recurrentis, angestrebt werden.
Im Rahmen dieser Arbeit wurde die schnelle Energietransfer- (EET) und Elektronentransfer (ET)-Dynamik unterschiedlichster Quantenpunkte (QD) spektroskopisch untersucht. Die untersuchten Systeme bestanden in den meisten Fällen aus Donor-Akzeptor-Paaren, bei denen die Halbleiternanokristalle als Donor fungierten. Der Fokus lag dabei auf der gezielten Anpassung des Donors, um die optimale Funktionalität zu erreichen. Die Untersuchung der Nanokristalle erstreckte sich daher von einfachen Kernen über verschiedene Kern-Schale-Partikel bis hin zu völlig anderen Strukturen wie Nanoplatelets (NPL). Als Akzeptor wurden eine Vielzahl von Molekülen verwendet, die sich als Elektronen- und/oder Energieakzeptoren für die verschiedenen QDs eignen.
Eine überlebenswichtige Eigenschaft von Mensch und Tier ist es, sich bei Gefahr durch eine Schreckreaktion in Sicherheit zu bringen. Doch woran erkennt ein Organismus, in welcher Situation es „sinnvoll“ wäre, sich zu erschrecken und welche Eigenschaften sensorischer Stimuli tragen zu dem Gefahreneindruck bei? Bei plötzlich eintretenden, lauten auditorischen Reizen kann es zur Auslösung der akustischen Schreckreaktion kommen. Dies führt bei Menschen, aber auch bei kleineren Säugetieren zu einer reflexartigen Kontraktion der Nacken-, Gesichts- und Skelettmuskulatur. Die Erforschung der akustisch evozierten Schreckreaktion (ASR) dient dem besseren Verständnis der neurobiologischen Grundlagen sensorischer Verarbeitung. Modulationen der ASR mithilfe von Präpulsen (Präpulsinhibition) ermöglichen Einblicke in die Funktion der Kochlea, des Hörnervs, der Hirnstammstrukturen und anderer beteiligter Gehirnregionen.
In dieser Arbeit wurden kurzzeitige Änderungen von Frequenz oder Intensität des akustischen Hintergrundes als neuartige Präpulse untersucht. Die Bedeutung verschiedener Reizparameter dieser Präpulse wurde in der vorliegenden Arbeit zum ersten Mal systematisch erforscht. Um zu prüfen, welche Präpulsstimulationen eine Inhibition der ASR auslösen können, wurde eine Reihe von Parametern umfassend getestet. In einem weiteren Schritt wurde analysiert, ob es mithilfe von gezielten Änderungen von Frequenz oder Intensität möglich sein könnte, Unterscheidungsschwellen, oder gar Hörschwellen von Versuchstieren zu bestimmen.
Die Experimente zur Modulation der ASR wurden mit weiblichen Sprague Dawley-Ratten durchgeführt. Dabei wurde eine Vielzahl von Verhaltensparadigmen untersucht. Dazu zählten Präpulse mit unterschiedlichem Frequenzgehalt und variabler Dauer. Zusätzlich wurden neuartige Paradigmen etabliert, um die Fähigkeit zur Frequenz- und Intensitätsdiskriminierung zu untersuchen. Hierbei wurde der Frequenzgehalt oder die Intensität einer kontinuierlichen Hintergrundstimulation verändert, um eine Präpulswirkung zu erzeugen. Um die Möglichkeiten der Bestimmung von Hörschwellen mittels der Präpulsinhibition (PPI) zu ergründen, wurde die Intensität von Präpulsen systematisch verändert. Die so generierten Schwellenwerte wurden durch die Messung früher akustisch evozierter Hirnstammpotenziale verifiziert. Schließlich sollten, unter Zuhilfenahme der Signaldetektionstheorie, aus den erhobenen Daten diverse Schwellen bestimmt werden: Für die Intensitätsänderungen der Präpulse in Stille wurden Hörschwellen bestimmt, während bei Änderungen der Frequenz und Intensität Unterscheidungsschwellen bestimmt werden sollten.
Mit steigender Größe eines Frequenzsprungs in einer kontinuierlichen Hintergrundstimulation war eine stärkere Inhibition der ASR feststellbar; ein Effekt, der stark von der Hintergrundfrequenz abhängig war. Bei einer Stimulation mit 8 kHz konnten signifikant höhere Inhibitionswerte erzielt werden als mit 16 kHz. Bei der Untersuchung des Zeitablaufs der Stimulation ergab sich, dass eine abgesetzte Stimulation mit einer Abweichung von 80 ms Dauer bis 50 ms vor dem Schreckreiz für die höchsten Inhibitionen sorgte.
Die durch eine Intensitätsänderung einer kontinuierlichen Hintergrundstimulation ausgelöste PPI hing primär von der Größe und Richtung des Intensitätssprungs ab. Mit zunehmender Sprunggröße stiegen die Inhibitionswerte an. Eine Erhöhung der Hintergrundintensität um 10 dB hatte einen signifikanten Einfluss auf die Inhibitionswerte. Auch hier zeigte sich eine höhere Sensitivität in Form von höheren Inhibitionen für Stimuli mit einer Hintergrundfrequenz von 8 kHz als für alle anderen getesteten Hintergrundfrequenzen.
Die Bestimmung von Hörschwellen mittels intensitätsabhängiger PPI wies im Vergleich mit den elektrophysiologisch bestimmten Hörschwellen ein heterogenes Bild mit starken individuellen Schwankungen auf: Bei etwa der Hälfte der Tiere waren die Hörschwellen beider Messungen sehr vergleichbar, bei den übrigen Tieren konnten mittels PPI für eine oder mehrere Frequenzen keine aussagekräftigen Hörschwellen erzielt werden. Die elektrophysiologisch bestimmten Hörschwellen waren am sensitivsten, während PPI-Stimulationen signifikant höher waren. Außerdem bewirkten PPI-Stimulationen mit Reintönen signifikant sensitivere Hörschwellen im Vergleich zu einem Schmalbandrauschen.
Für die Bestimmung der Unterscheidungsschwellen von Frequenzänderungen konnte beobachtet werden, dass die Tiere auf Frequenzsprünge hin zu niedrigeren Frequenzen signifikant sensibler reagierten, als hin zu Aufwärtssprüngen (-1.2 bzw. +4.5%). Bei der Intensitätsunterscheidung hingegen konnte beobachtet werden, dass die Tiere signifikant sensitiver auf Intensitätserhöhungen als auf Erniedrigungen reagierten (-5.9 bzw. +2.7 dB).
Zusammenfassend konnte in der vorliegenden Arbeit festgestellt werden, dass die PPI zur Bestimmung von absoluten Hörschwellen starken Schwankungen unterlag, sodass diese Methode nur eingeschränkt als Alternative zu operanter Konditionierung oder elektrophysiologischen Ableitungen in Frage kommt. Des Weiteren erzeugten bereits kleine Änderungen des Frequenzgehalts oder der Intensität einer Hintergrundstimulation eine robuste PPI. Somit können reflexbasierte Messungen mit überschwelligen Stimuli genutzt werden, um Unterscheidungsschwellen in Versuchstieren zu bestimmen. Diese Herangehensweise stellt also eine vielversprechende Methode dar, um Hörstörungen zu untersuchen, die nach einem Schalltrauma auftreten können. In einem nächsten Schritt könnte sie zur weiteren Charakterisierung von verstecktem Hörverlust beitragen.
Nanoarzneimittel haben in den letzten Jahren in der Therapie verschiedener Erkrankungen immer mehr an Bedeutung gewonnen. Dadurch hat auch die Anzahl zugelassener Arzneimittel mit an Arzneistoffträgern wie Liposomen gebundenen Wirkstoffen zugenommen. Weil für die Zulassung, neben der Wirksamkeit und Unbedenklichkeit, auch die Qualität der neuen Arzneimittel gewährleistet sein muss, spielen die verschiedenen Eigenschaften der Arzneistoffträger eine wichtige Rolle in der Qualitätskontrolle. Neben der Partikelgröße, der Partikelgrößenverteilung und der Oberflächenladung spielt die (Rest-)Kristallinität des Wirkstoffs und die Wirkstofffreisetzung eine wesentliche Rolle für die erfolgreiche in vivo-Performance von Nanoarzneimitteln. Zur Bestimmung der Wirkstofffreisetzung aus kolloidalen Arzneistoffträgern wie Liposomen, Nanopartikeln oder Mizellen gibt es bis heute keine Standardmethoden. In der Forschung und der pharmazeutischen Industrie werden folglich verschiedene Methoden wie Filtration, Zentrifugation oder Dialyse verwendet, um den freigesetzten Wirkstoff zu bestimmen. Dabei ist die Wahl der Separationsmethode auf die Eigenschaften der Arzneistoffträger abzustimmen.
In der vorliegenden Arbeit wurde eine dialysebasierte Apparatur, der Dispersion Releaser (DR), zur Untersuchung der in vitro Wirkstofffreisetzung aus kolloidalen Trägersystemen eingesetzt. Diese kann direkt mit den Apparaturen I/II der Arzneibücher der Europäischen Union (Ph. Eur.) und der Vereinigten Staaten (USP) gekoppelt werden. Zur Untersuchung der Wirkstofffreisetzung wird die Formulierung in das Donorkompartiment gegeben, sodass der freigesetzte Wirkstoff infolge über die Dialysemembran in das Akzeptorkompartiment permeiert. Dort kann dieser mittels HPLC analysiert werden. Besonders hervorzuheben ist das synchrone Rühren in beiden Kompartimenten des DR, worüber andere dialysebasierte Apparaturen nicht verfügen.
Die Entwicklung und Patentierung eines funktionsfähigen Prototyps des DR erfolgte an der Goethe Universität, Frankfurt am Main und wurde im Rahmen dieser Arbeit gemeinsam mit der Pharma Test Apparatebau AG (Hainburg, Deutschland) zu einer kommerziell erwerbbaren Apparatur (Pharma Test Dispersion Releaser, PTDR) weiterentwickelt. Innerhalb dieser Kollaboration wurde der Prototyp des DR unter Einbezug der Anforderungen der pharmazeutischen Industrie rekonstruiert. Eine erleichterte Anwendung für den Nutzer wurde dabei mitberücksichtigt.
Die finale Apparatur wurde zuletzt einer ausgiebigen Validierung unterzogen, bei der Diclofenac und Hydrocortison als Modellarzneistoffe dienten. Neben Untersuchungen zur Hydrodynamik und dem Einfluss der Umdrehungszahl auf die Membranpermeationsrate kM wurde eine Methode mit Gold-Nanopartikeln zur Bestimmung der Dichtigkeit des Systems entwickelt. Hierbei wurden Messungen mit einer UV/Vis-Methode und mit dynamischer Lichtstreuung durchgeführt, um die Abwesenheit der Goldpartikel im Akzeptorkompartiment nachzuweisen. Der Einfluss von Proteinen im Freisetzungsmedium auf die Membran-permeation wurde ebenfalls untersucht.
Der DR wurde ursprünglich zur Untersuchung von parenteralen Nanoformulierungen entwickelt. Aufgrund der bisher noch nicht erfolgten Untersuchung von halbfesten Zubereitungen im DR, wurde die Apparatur im Rahmen dieser Forschungsarbeit für zwei verschiedene Diclofenac-Gele (Voltaren® Emulgel, Olfen® Gel) unter verschiedenen Bedingungen evaluiert. Dabei konnte unter non-sink-Bedingungen der Einfluss der lipophilen Phase des Voltaren® Emulgels (GlaxoSmithKline Consumer Healthcare GmbH & Co. KG, München, Deutschland) gezeigt werden. Im Vergleich zum fettfreien Olfen® Gel (Mepha Pharma AG, Basel, Schweiz) zeigte Voltaren® Emulgel eine vollständige Freisetzung unter den erschwerten Löslichkeitsbedingungen.
Mit Hydrocortison als Modellsubstanz wurden vier verschiedene Proliposomen zur vaginalen An¬wendung formuliert. Neben der Charakterisierung der Partikelgröße und der Verkapselungs¬effizienz wurden Messungen mit dynamischer Differenzkalorimetrie durch-geführt und Aufnahmen zur morphologischen Charakterisierung mittels Transmissions-elektronen¬mikroskopie der Liposomen erstellt. Die Wirkstofffreisetzung des Hydrocortisons aus dem rekonstituierten liposomalen Gel sowie die Permeabilität über eine Zellmonoschicht wurde vergleichend untersucht. Dabei wurden Zelllinien aus humanem Cervixkarzinom beziehungsweise Endometriumkarzinom eingesetzt. Die Unterschiede der Formulierungen konnten vom DR sensitiver erfasst werden und die Verkapselungseffizienz als relevanter Faktor für die in vivo-Performance festgelegt werden.
Weil die tatsächliche Wirkstofffreisetzung durch die Permeation über die Dialysemembran überlagert werden kann, wurde neben der Standardisierung der Konstruktion die Auswertung mit Hilfe eines neuen mathematischen Modells, das auf dem Fick’schen Diffusionsgesetz basiert, verbessert. Das Normalisieren des Freisetzungsprofils mit Hilfe des mathematischen Modells dient dazu, die tatsächliche Wirkstofffreisetzung zu berechnen und den Vergleich verschiedener Freisetzungen ohne den Einfluss der Membranpermeation zu ermöglichen. Im Zuge der Validierung des DR wurde das mathematische Modell ebenfalls erfolgreich validiert.
In der vorliegenden Forschungsarbeit wurde eine neue Konstruktion des DR für die kommerzielle Anwendung entwickelt und validiert. Nebenbei wurde der Auswerteprozess zur Berechnung der diffusionsbereinigten Wirkstofffreisetzung vereinheitlicht und validiert. Zuletzt wurde das Anwendungsgebiet des DR von parenteralen Nanoformulierungen auf halbfeste Arzneiformen erweitert.
This chapter examines Edmond Jabès, who chose to write his oeuvre in French despite his Jewish-Arabic origins and his being conversant in both Hebrew and Arabic. French was never a true 'mother tongue' to him but rather 'a foreign one'. This poetical choice was also instrumental to his creation of a cosmos that is very clearly defined by 'la page blanche', or the 'blank page'. His writing develops this idea, both literally and metaphorically. A blank sheet is the only thing a writer has to work with at the start of every writing act, therefore it represents a kind of material opposition that all writers must overcome. It represents in this context an existential nothingness that precedes and simultaneously escapes both human and divine creation. In Jabès's writings, a blank page has two connotations at once: a condition for writing and nothingness. This ambivalent condition results in the paradoxical assumption that his 'mother tongue is a foreign language', because it cannot offer the same spiritual intimacy as another language, say, the Holy Language, and because the writer's 'mother tongue' - and, by extension, human language - is always impure and infiltrated by foreignness.
Dual coding theories of knowledge suggest that meaning is represented in the brain by a double code, which comprises language-derived representations in the Anterior Temporal Lobe and sensory-derived representations in perceptual and motor regions. This approach predicts that concrete semantic features should activate both codes, whereas abstract features rely exclusively on the linguistic code. Using magnetoencephalography (MEG), we adopted a temporally resolved multiple regression approach to identify the contribution of abstract and concrete semantic predictors to the underlying brain signal. Results evidenced early involvement of anterior-temporal and inferior-frontal brain areas in both abstract and concrete semantic information encoding. At later stages, occipito-temporal regions showed greater responses to concrete compared to abstract features. The present findings shed new light on the temporal dynamics of abstract and concrete semantic representations in the brain and suggest that the concreteness of words processed first with a transmodal/linguistic code, housed in frontotemporal brain systems, and only after with an imagistic/sensorimotor code in perceptual and motor regions.
We present a massively parallel framework for computing tropicalizations of algebraic varieties which can make use of symmetries using the workflow management system GPI-Space and the computer algebra system Singular. We determine the tropical Grassmannian TGr0(3,8). Our implementation works efficiently on up to 840 cores, computing the 14763 orbits of maximal cones under the canonical S8-action in about 20 minutes. Relying on our result, we show that the Gröbner structure of TGr0(3,8) refines the 16-dimensional skeleton of the coarsest fan structure of the Dressian Dr(3,8), except for 23 orbits of special cones, for which we construct explicit obstructions to the realizability of their tropical linear spaces. Moreover, we propose algorithms for identifying maximal-dimensional cones which belong to positive tropicalizations of algebraic varieties. We compute the positive Grassmannian TGr+(3,8) and compare it to the cluster complex of the classical Grassmannian Gr(3,8).
A new fossil ceratopogonid genus and species from Lower Cretaceous Lebanese amber, Baskintoconops maaloufi Pielowska-Ceranowska gen. et sp. nov., is described and illustrated. The studied material originates from a newly discovered amber site in the Lebanese village Baskinta at a locality dubbed Qanat Bakish. The described genus is typified by its wing venation pattern combining characters of genera Fossileptoconops and Jordanoconops belonging to the subfamily Leptoconopinae.
Measurements of inclusive charged-particle jet production in pp and p-Pb collisions at center-of-mass energy per nucleon-nucleon collision sNN−−−√=5.02 TeV and the corresponding nuclear modification factor RchjetpPb are presented, using data collected with the ALICE detector at the LHC. Jets are reconstructed in the central rapidity region |ηjet|<0.5 from charged particles using the anti-kT algorithm with resolution parameters R=0.2, 0.3, and 0.4. The pT-differential inclusive production cross section of charged-particle jets, as well as the corresponding cross-section ratios, are reported for pp and p-Pb collisions in the transverse momentum range 10<pchT,jet<140 GeV/c and 10<pchT,jet<160 GeV/c, respectively, together with the nuclear modification factor RchjetpPb in the range 10<pchT,jet<140 GeV/c. The analysis extends the pT range of the previously-reported charged-particle jet measurements by the ALICE Collaboration. The nuclear modification factor is found to be consistent with one and independent of the jet resolution parameter with the improved precision of this study, indicating that the possible influence of cold nuclear matter effects on the production cross section of charged-particle jets in p-Pb collisions at sNN−−−√=5.02 TeV is smaller than the current precision. The obtained results are in agreement with other minimum bias jet measurements available for RHIC and LHC energies, and are well reproduced by the NLO perturbative QCD POWHEG calculations with parton shower provided by PYTHIA8 as well as by JETSCAPE simulations.
Im Rahmen des von der KfW Stiftung geförderten Artenschutzprojektes „Erhaltungskulturen von bedrohten Pflanzen in Hessen“ wurden in den Jahren 2014–2018 von 15 gefährdeten Pflanzenarten in verschiedenen Regionen Hessens Samen gesammelt und im Botanischen Garten der Stadt Frankfurt am Main vermehrt. In Zusammenarbeit mit Naturschutzbehörden, Forstämtern und Naturschutzverbänden wurden dokumentierte Wiederansiedlungsmaßnahmen oder Populationsstärkungen durchgeführt. Überzähliges Saatgut wurde im Botanischen Garten eingefroren und kann für zukünftige Artenschutzprojekte verwendet werden. Bei den 15 gefährdeten Arten handelt es sich um Allium strictum, Festuca albensis, Fumana procumbens, Iris spuria, Jurinea cyanoides, Mibora minima, Moenchia erecta, Nigella arvensis, Poa badensis, Scleranthus verticillatus, Sedum villosum, Spergula pentandra, Ventenata dubia, Veronica acinifolia und Vicia orobus. Alle diese Arten sind als Verantwortungsarten Bestandteil der hessischen Biodiversitätsstrategie. Im Jahr 2021, also drei Jahre nach dem Ende des Projektes, ergab eine Erfolgskontrolle folgendes Ergebnis: Von insgesamt 50 Wiederansiedlungsversuchen an 34 Orten sind immerhin 25 (50 %) erfolgreich. Weitere 5 (10 %) sind möglicherweise ebenfalls gelungen, jedoch sind die neu begründeten Populationen sehr klein. Eindeutig misslungen sind 20 (40 %). Die Gründe für das Misslingen sind meist klar erkennbar und werden thematisiert: Manchmal war der Zielort ungünstig gewählt, oder die verwendete Ansiedlungsmethode erwies sich als ungeeignet. Das Projekt konnte dazu beitragen, unser Wissen über Ökologie und Verbreitung sowie Gefährdung und Schutzbedarf dieser Verantwortungsarten teils erheblich zu vermehren. Damit leistet es auch einen Beitrag zum Schutz der bereits bestehenden Altpopulationen.
Campanula cervicaria steht in Hessen kurz vor dem Aussterben und wurde im Rahmen des Kooperationsvertrages zwischen der Botanischen Vereinigung für Naturschutz in Hessen (BVNH) und dem Land Hessen untersucht. Das Ziel der Untersuchung war, herauszufinden wo Campanula cervicaria in Hessen noch vorkommt und wie groß die Populationen sind. In Hessen sind aktuell noch 20 Populationen vorhanden. Fast 90 % der Populationen befinden sich in der Umgebung von Lißberg, Hirzenhain und Gedern im Westlichen Unteren Vogelsberg. Zwei weitere liegen bei Jossa im Spessart und bei Ober-Gleen im Nördlichen Vogelsberg-Vorland. Die größte Population befindet sich an der Kreisstraße 216 bei Hirzenhain am Forsthaus Luisenlust. Campanula cervicaria wächst in Hessen an sonnenexponierten Wegböschungen und Windwurfflächen, meist zusammen mit Hypericum hirsutum und Brachypodium sylvaticum. Um auch zukünftig die Bestände von Campanula cervicaria zu sichern, müsste ein Artenhilfskonzept erarbeitet werden.
Betrachtet man die Anzahl der in Hessen ausgestorbenen Pflanzenarten über den gesamten Zeitraum ab Beginn der floristischen Erforschung, so scheint ein Zusammenhang zwischen der geringeren Anzahl ausgestorbener Arten in den letzten Dekaden und den gleichzeitig vermehrt ausgewiesenen Naturschutzgebieten zu bestehen. Es stellt sich daher die Frage, ob es auch einen kausalen Zusammenhang gibt, oder ob andere Faktoren einen Einfluss auf das Ausmaß an ausgestorbenen Pflanzenarten haben. Für die Entwicklung der Aussterberate in den kommenden Jahrzehnten wird es nötig werden, alle Vorkommen der derzeit vom Aussterben bedrohten Pflanzenarten dahingehend zu überprüfen, ob ihr Bestand gesichert und erhalten werden kann. So kann dann die Frage beantwortet werden, ob es gelungen ist, das Aussterben von Pflanzenarten in Hessen zu stoppen oder lediglich zu verlangsamen.

Unsere bisherige Kenntnis zur Situation von Crepis praemorsa (Abbiss-Pippau) war ungenügend und wurde im Rahmen eines Kooperationsprojektes mit dem Hessischen Landesamt für Naturschutz, Umwelt und Geologie (HLNUG) erheblich verbessert. Die Untersuchungen zeigen, dass es in Hessen offenbar nur noch acht Vorkommen dieser Art gibt: Drei sehr kleine Reliktpopulationen an der Bergstraße, eine im nordhessischen Bergland und vier in der Rhön, davon zwei sehr kleine. Keine der Populationen kann als groß bezeichnet werden, die Anzahl der Individuen liegt zwischen wenigen Exemplaren bis zum niedrigen dreistelligen Bereich. Lediglich eine Population erscheint aktuell ungefährdet. Fehlende Nutzung oder Pflege und in der Folge Verbrachung und Verbuschung sowie Lichtmangel durch Beschattung sind maßgebliche Gefährdungen. Ohne aktive Hilfsmaßnahmen werden vier dieser acht Populationen in naher Zukunft erlöschen.

Proteins often bind with each other forming complexes to fulfill their function. Such protein complexes assemble along an ordered pathway. An assembled protein complex can often be divided into structural and functional modules. Knowing the order of assembly and the modules of a protein complex is important to understand biological processes and treat diseases related to misassembly.
Typical structures of the Protein Data Bank (PDB) contain two to three subunits and a few thousand atoms. Recent developments have led to large protein complexes being resolved. The increasing number and size of the protein complexes demand for computational assistance for the visualization and analysis. One such large protein complex is respiratory complex I accounting for 45 subunits in Homo sapiens.
Complex I is a well understood protein complex that served as case study to validate our methods.
Our aim was to analyze time-resolved Molecular Dynamics (MD) simulation data, identify modules of a protein complex and generate hypotheses for the assembly pathway of a protein complex. For that purpose, we abstracted the topology of protein complexes to Complex Graphs of the Protein Topology Graph Library (PTGL). The subunits are represented as vertices, and spatial contacts as edges. The edges are weighted with the number of contacts based on a distance threshold. This allowed us to apply graph-theoretic methods to visualize and analyze protein complexes.
We extended the implementations of two methods to achieve a computation of Complex Graphs in feasible runtimes. The first method skipped checks for contacts using the information which residues are sequential neighbors. We extended the method to protein complexes and structures containing ligands. The second method introduced spheres encompassing all atoms of a subunit and skipped the check for contacts if the corresponding spheres do not overlap. Both methods combined allowed skipping up to 93 % of the checks for contacts for sample complexes of 40 subunits compared to up to 10 % of the previous implementation. We showed that the runtime of the combined method scaled linearly with the number of atoms compared to a non-linear scaling of the previous implementation We implemented a third method fixing the assignment of an orientation to secondary structure elements. We placed a three-dimensional vector in each secondary structure element and computed the angle between secondary structure elements to assign an orientation. This method sped up the runtime especially for large structures, such as the capsid of human immunodeficiency virus, for which the runtime decreased from 43 to less than 9 hours.
The feasible runtimes allowed us to investigate two data sets of MD trajectories of respiratory complex I of Thermus thermophilus that we received. The data sets differ only by whether ubiquinone is bound to the complex. We implemented a pipeline, PTGLdynamics, to compute the contacts and Complex Graphs for all time steps of the trajectories. We investigated different methods to track changes of contacts during the simulation and created a heat map put onto the three-dimensional structure visualizing the changes. We also created line plots to visualize the changes of contacts over the course of the simulation. Both visualizations helped spotting outstandingly flexible or rigid regions of the structure or time points of the simulation in which major dynamics occur.
We introduced normalizations of the edge weights of Complex Graphs for identi-fying modules and predicting the assembly pathway. The idea is to normalize the number of contacts for the number of residues of a subunit. We defined five different normalizations.
To identify structural and functional modules, we applied the Leiden graph clustering algorithm to the Complex Graphs of respiratory complex I and the respiratory supercomplex. We examined the results for the different normalizations of the weights of the Complex Graphs. The absolute edge weight produced the best result identifying three of four modules that have been defined in the literature for respiratory complex I.
We applied agglomerative hierarchical clustering to the edges of a Complex Graph to create hypotheses of the assembly pathway. The rationale was that subunits with an extensive interface in the final structure assemble early. We tested our method against two existing methods on a data set of 21 proteins with reported assembly pathways. Our prediction outperformed the other methods and ran in feasible runtimes of a few minutes at most.
We also tested our method on respiratory complex I, the respiratory supercomplex and the respiratory megacomplex. We compared the results for the different normalizations with an assembly pathway of respiratory complex I described in the literature. We transformed the assembly pathways to dendrograms and compared the predictions to the reference using the Robinson-Foulds distance and clustering information distance. We analyzed the landscape of the clustering information distance by generating random dendrograms and showed that our result is far better than expected at random. We showed in a detailed analysis that the assembly prediction using one normalization was able to capture key features of the assembly pathway that has been proposed in the literature.
In conclusion, we presented different applications of graph theory to automatically analyze the topology of protein complexes. Our programs run in feasible runtimes even for large complexes. We showed that graph-theoretic modeling of the protein structure can be used to analyze MD simulation data, identify modules of protein complexes and predict assembly pathways.
SpikeShip: a method for fast, unsupervised discovery of high-dimensional neural spiking patterns

Neural coding and memory formation depend on temporal spiking sequences that span high-dimensional neural ensembles. The unsupervised discovery and characterization of these spiking sequences requires a suitable dissimilarity measure to spiking patterns, which can then be used for clustering and decoding. Here, we present a new dissimilarity measure based on optimal transport theory called SpikeShip, which compares multi-neuron spiking patterns based on all the relative spike-timing relationships among neurons. SpikeShip computes the optimal transport cost to make all the relative spike timing relationships (across neurons) identical between two spiking patterns. We show that this transport cost can be decomposed into a temporal rigid translation term, which captures global latency shifts, and a vector of neuron-specific transport flows, which reflect inter-neuronal spike timing differences. SpikeShip can be effectively computed for high-dimensional neuronal ensembles, has a low (linear) computational cost that has the same order as the spike count, and is sensitive to higher-order correlations. Furthermore SpikeShip is binless, can handle any form of spike time distributions, is not affected by firing rate fluctuations, can detect patterns with a low signal-to-noise ratio, and can be effectively combined with a sliding window approach. We compare the advantages and differences between SpikeShip and other measures like SPIKE and Victor-P urpura distance. We applied SpikeShip to large-scale Neuropixel recordings during spontaneous activity and visual encoding. We show that high-dimensional spiking sequences detected via SpikeShip reliably distinguish between different natural images and different behavioral states. These spiking sequences carried complementary information to conventional firing rate codes. SpikeShip opens new avenues for studying neural coding and memory consolidation by rapid and unsupervised detection of temporal spiking patterns in high-dimensional neural ensembles.
Anticipating future events is a key computational task for neuronal networks. Experimental evidence suggests that reliable temporal sequences in neural activity play a functional role in the association and anticipation of events in time. However, how neurons can differentiate and anticipate multiple spike sequences remains largely unknown. We implement a learning rule based on predictive processing, where neurons exclusively fire for the initial, unpredictable inputs in a spiking sequence, leading to an efficient representation with reduced post-synaptic firing. Combining this mechanism with inhibitory feedback leads to sparse firing in the network, enabling neurons to selectively anticipate different sequences in the input. We demonstrate that intermediate levels of inhibition are optimal to decorrelate neuronal activity and to enable the prediction of future inputs. Notably, each sequence is independently encoded in the sparse, anticipatory firing of the network. Overall, our results demonstrate that the interplay of self-supervised predictive learning rules and inhibitory feedback enables fast and efficient classification of different input sequences.
The differential cross section for Z0 production, measured as a function of the boson's transverse momentum (pT), provides important constraints on the evolution of the transverse momentum dependent parton distribution functions (TMDs). The transverse single spin asymmetry (TSSA) of the Z0 is sensitive to one of the polarized TMDs, the Sivers function, which is predicted to have the opposite sign in p+p →W/Z+X from that which enters in semi-inclusive deep inelastic scattering. In this Letter, the STAR Collaboration reports the first measurement of the Z0/γ∗ differential cross section as a function of its pT in p+p collisions at a center-of-mass energy of 510 GeV, together with the Z0/γ∗ total cross section. We also report the measurement of Z0/γ∗ TSSA in transversely polarized p+p collisions at 510 GeV.
The nitidus species group of the bee genus Scrapter Lepeletier & Serville, 1828 is redefi ned, revised and 15 species are described as new for science: S.caeruleus sp. nov. ♀, S.confusus sp. nov. ♀♂, S.convexoides sp. nov. ♂, S. convexus sp. nov. ♀♂, S. crassipunctatus sp. nov. ♀♂, S.felicis sp. nov. ♀♂, S.fl avipunctatus sp. nov. ♀♂, S.imparilis sp. nov. ♀♂, S. littoralis sp. nov. ♀, S.longicornis sp. nov. ♂, S.montanus sp. nov. ♀♂, S.mpumalangensis sp. nov. ♀♂, S.obtusus sp. nov. ♀♂, S. perpunctatulus sp. nov. ♂ and S.variabilis sp. nov. ♀♂. The previously unknown males of S. divergens (Friese, 1925), S. semirufus Cockerell, 1932 and S. perpunctatus Cockerell, 1933 are described for the fi rst time. All currently known 28 species of the S. nitidus species group are redescribed, imaged and included in a key to facilitate their identifi cation.
Bumblebees (Bombus Latreille, 1802), because of their large body size, bright colours and activity at times and places that coincide with biologists, are an example of a group of insects that is particularly well represented in museum collections. This is important if taxonomic revisions are to achieve greater comparability among species. Bumblebees have also attracted particular attention because they are especially ecologically and economically valuable for pollination in north temperate regions, where they are now becoming increasingly threatened. I argue that the what, the where, and the how of effective conservation management may be informed by understanding the divergent characteristics that have affected their biogeographical past: by helping us to see ‘the woods’, not just ‘the trees’, of their habitat needs. Identifying suitable habitat should be part of reconstructing historical biogeography within taxonomic revisions. For bumblebees, for example, biogeographical analysis associates major taxonomic groups either with flower-rich lowland grasslands or with flower-rich montane grasslands, highlighting their contrasting requirements for: nest sites, flowers of different depths, pollen-plant families, and especially the differing importance of early spring and late summer flowers for breeding success. This broad view of species groups helps filter the less important idiosyncrasies from local case studies in order to focus conservation actions.
Bostrichidae (Coleoptera) are a family of beetles with specialized xylophagous feeding habits that allow consumption of dry woody plant tissues. Bostrichids are often polyphagous and therefore pose a significant threat to many agricultural and forestry products, particularly in tropical regions. Bostrichids are commonly detected in solid wood packaging material at in ternational ports of entry. Notably, Sinoxylon anale Lesne has been intercepted in wood crates and pallets worldwide and has now become established in Brazil. This paper reports the first documented establishment of S. anale in Brazil, being found both in domestic wood pallets and within native forest and monoculture. The origin of these populations remains uncertain, but introductions through infested wood packaging at ports of entry is a likely scenario. Similarly, the exact time of establishment is unknown. Given that S. anale adults are attracted to light and ethanol, trapping using light or ethanol could be used in monitoring surveys. This species typically infests dead or decaying woody material, and therefore does not pose a direct threat to healthy, live trees. Although more common in tropical regions, S. anale has exhibited some adaptability to temperate climates, which may allow it to spread across the tropical and subtropical regions of Brazil and potentially to other parts of South America.

Firms, researchers, and policy makers often want to measure consumption and especially how events, promotions, or policies affect it. Measuring consumption reactions is often hard. Firms lack access to competitors’ sales data and regularly do not share their own with outsiders. Large samples of smartphone location data could solve this problem. This article describes a research project using smartphone location data to estimate consumption reactions to political conflict during the Trump presidency.

Sie wollen mehr Aufmerksamkeit für den Biodiversitätshotspot Totholz schaffen. Für ihre Idee, Totholzinseln im Frankfurter Stadtgebiet anzulegen, wurden sie kürzlich mit dem dritten Platz des Ideenwettbewerbs Biodiversität von Goethe-Universität, Palmengarten, der Senckenberg Gesellschaft für Naturforschung, dem Dezernat für Klima, Umwelt und Frauen der Stadt Frankfurt und der Frankfurter Sparkasse ausgezeichnet.
Lysosomes are major degradative organelles that contain enzymes capable of breaking down proteins, nucleic acids, carbohydrates, and lipids. In the last decade, new discoveries have traced also important roles for lysosomes as signalling hubs, affecting metabolism, autophagy and pathogenic infections. Therefore, maintenance of a healthy lysosome population is of utmost importance to the cell to respond to both stress conditions and also homeostatic signalling. For example, for minor perturbations to the lysosomal membrane, the cell activates repair processes which seal membrane nicks. For more extensive damage, autophagy is activated to remove damaged organelles from the cell. on the other hand, during pathogen invasion host cells have also evolved mechanisms to hijack the endolysosomal pathway to facilitate their own growth and replication in host cells.
The first part of the thesis work focuses on a lysosomal regeneration program which is activated under conditions where the entire lysosomal pool of the cell is damaged. Upon extensive membrane damage induced by the lysosomotropic drug LLOMe, the cell activates a regeneration pathway which helps in the formation of new functional lysosomes by recycling damaged membranes. I have identified the molecules important for this novel pathway of lysosomal regeneration and showed how the protein TBC1D15 orchestrates this process to regenerate functional organelles from completely damaged membrane masses in the first 2 hours following lysosomal membrane damage. This process resembles the process of auto- lysosomal reformation (ALR)- involving the formation of lysosomal tubules which are extended along microtubules and cleaved in a dynamin2 dependent manner to form proto-lysosomes which develop into fully functional mature lysosomes. These lysosomal tubules are closely associated with ATG8 positive autophagosomal membranes and require ATG8 proteins to bind to the lysophagy receptor LIMP2 on damaged membranes. This process is physiologically important under conditions of crystal nephropathy where calcium oxalate crystals induce damage to lysosomal membranes in nephrons in kidney disease.
The second part of the thesis shows how the endolysosomal system of the cell is hijacked by the bacteriaLegionella pneumophila. During Legionella infection the formation of conventional ATG8 positive autophagosomes are blocked due to the protease activity of the bacterial effector protein RavZ which cleaves lipidated ATG8 proteins from autophagosomal membranes. The SidE effectors of Legionella modify STX17 and SNAP29 by the process of non-canonical ubiquitination called phosphoribose-linked serine ubiquitination (PR-Ub). These proteins are essential for the formation of the autophagosomal SNARE complex which is used for fusion of the autophagosome with the lysosome. Upon Legionella infection, PR-UB of STX17 aids in formation of autophagosome-like replication vacuoles. ThesevacuolesdonotfusewiththelysosomebecauseSNAP29isalsoPR-Ubmodified. PR-UbofSTX17 and SNAP29 sterically blocks the formation of the autophagosomal-SNARE complex thereby preventing fusion of the autophagosome with the lysosome. As a result, Legionella can replicate in autophagosome- like vacuoles which do not undergo lysosomal degradation. In absence of PR-Ub modified STX17, bacterial replication is compromised when measured by bacterial replication assays in lung epithelial (A549) cells.
Taken together, this thesis highlights two important aspects of the autophagy-lysosomal system- how it responds to extensive membrane damage and its importance in Legionella pneumophila infection. Extensive damage to lysosomal membranes triggers a rapid regeneration process to partially restore lysosomal function before the effects of TFEB dependent lysosomal biogenesis becomes apparent. On the other hand, Legionella pneumophila infection segregates the lysosomes from the rest of the endo-lysosomal system by blocking autophagosome-lysosome fusion. Though lysosomes remain active, they are incapable of degrading pathogens since pathogen containing vacuoles do not fuse with the lysosome.
We study the production of the light JPC=1−+ hybrid isoscalars η1′(1855) and the yet undiscovered η1(1660) as intermediate states in the radiative decays of the charmonium (J/ψ) to two conventional mesons using a flavor symmetric Lagrangian. For this purpose, we use the J/ψ→γη1′(1855)→γηη′ process as the reference. We find that some of the decay channels have branching ratios similar to or larger than that of the γηη′ channel and are sensitive to the mixing between the hybrid isoscalars. We propose that relatively stable γηf1(1285) channel be explored for the presence of the light hybrid isoscalar η1hyb(1660). We also exploit the strong decay channels containing at least one vector meson to study the radiative decays of the whole hybrid nonet {π1(1600),K1hyb(1750),η1hyb(1660),η1′(1855)}. We find that the hybrids cannot radiatively decay into the I=0 pseudoscalars. Furthermore, the vector decay channels ((ρ/ω/ϕ)γ) of the hybrid isoscalars are sensitive to the strangeness content of the hybrids. We also provide estimates for the branching fractions for the radiative production and partial widths for the radiative decays of the hybrids.
N6-methyladenosine (m6A) is the most abundant and well understood modification in eukaryotic mRNA and was first identified in polyadenylated parts of the mRNA.The distinct distribution of m6A in the transcriptome with special enrichment in long internal exons, 39UTRs and around stop codons was uncovered by early biochemical work and later on antibody based sequencing techniques. The so called m6A writer, reader and eraser machinery is responsible for the dynamic and with that regulatory nature of the m6A modification. As m6A writer, the human N6-methyltransferase complex (MTC) cotranscriptionally methylates the central adenine within a RRACH (preferably GGACU) sequence context to form m6A in the nascent RNA chain.9–15 The catalytic core of the complex is formed by the two proteins METTL3 and METTL14, with the active site located in the methyltransferase domain (MTD) of METTL3.16–18 The DPPW motif near the methyl donor S-adenosylmethionine (SAM) binding site in this MTD was postulated to bind the target adenine during catalysis. Moreover, a positively charged groove in the METTL3-METTL14 interface, the C-terminal RGG domain in METTL14 and the zinc finger motifs in METTL3 were identified as important domains for RNA binding. However, to date there are no full-length or substrate-RNA-bound structures of the catalytic METTL3-METTL14 complex.
In addition, a set of accessory proteins assembles to the METTL3-METTL14 heterodimer to form the full MTC, mediated by WTAP that firmly binds to the N-terminal leader helix in METTL3.20 WTAP was shown to locate the whole complex to the nuclear speckles and can modulate m6A deposition to specific sites in the RNA. Moreover, WTAP acts as binding platform for other accessory proteins including VIRMA, RBM15, ZC3H13 and HAKAI that are mostly identified to mediate position specific methylation. For example, RBM15 was shown to mediates region-selective methylation in a WTAP dependent manner, directing specificity towards U-rich sequences.
The observed specificity of the methyltransferase complex to methylate only site specific DRACH sequenced is still poorly understood. Some possible modulators like the role of the accessory proteins are under investigation, however, the structural context of the RNA methylation sites or a structural preference of the complex have been mainly neglected so far. Moreover, the structural dynamics of this methylation process still remain elusive. This thesis contributes to the afore-mentioned aspects by analysis of the methylation process regarding RNA structure sensitivity with enzymatic activity assays and its dynamic nature by implementing a smFRET approach.
We hypothesized the target RNA secondary structure to be an additional important modulator of methylation efficiency, based on the RNA binding elements of the complex (positively charged binding groove, zinc finger domain, RGG domain) and the supposed target adenine binding in the active site. Here, we postulated the possibility for a flipped-out adenine to be of special relevance, which is closely related to the local stability of the target adenine containing structure. Moreover, efficient binding of the protein complex to the RNA should require the ability to anchor the RNA on both sides of the target sequence.
Chloritoid and kyanite coexist in metapelites from the high-pressure/low-temperature Massa Unit in the Alpi Apuane metamorphic complex (Northern Apennines, Italy). The composition of chloritoid is extremely variable throughout the Massa Unit. Fe-chloritoid occurs in association with hematite-free, graphite-bearing schists, whereas strongly zoned Fe-Mg chloritoid is found with hematite and kyanite. We investigated the effect of different bulk Fe2O3 contents in controlling chloritoid composition through phase equilibria modelling of four selected samples, representative of the different chloritoid-bearing parageneses found in the Massa Unit. The ferric iron content, measured through wet chemical titration, ranges from 0 (graphite-chloritoid schist) to 73% of the total iron (hematite-chloritoid schist). We show that Mg-rich chloritoid compositions and stability of kyanite at greenschist to blueschist facies conditions can be reproduced in the MnO–Na2O–K2O–FeO–MgO–Al2O3–SiO2–H2O–TiO2–O (MnNKFMASHTO) chemical system only considering the presence of significant amounts of ferric iron as part of the bulk composition. The stabilization of kyanite at lower grade is directly linked to the presence of Fe2O3, which renders the reactive bulk rock composition effectively enriched in Al2O3 with respect to Fe and Mg. We also document that high Fe2O3 contents exacerbate the effect of chloritoid fractionation, producing strongly zoned Fe-Mg-chloritoid grains. Finally, the P–T modelling of the Massa Units performed in this study allows, for the first time, the recognition of a two-stage evolution at peak conditions, with an earlier pressure peak (1.2–1.3 GPa at 350–400°C), and a later thermal peak (0.7–1.1 GPa at 440–480°C), compatible with subduction, underthrusting and exhumation of the Adria continental margin during growth of the Northern Apennine orogenic wedge.
Die vorliegende Dissertation mit dem Titel “Structural dynamics of eukaryotic H/ACA RNPs from Saccharomyces cerevisiae & Structural dynamics of the Guanidine-II riboswitch from Escherichia coli” besteht aus zwei Projekten. Das erste Projekt befasst sich mit den eukaryotischen H/ACA Ribonukleoproteinen (RNP) aus der Hefe. Diese können sequenzspezifisch in der RNA ein Uridin Nukleotid in das Rotationsisomer Pseudouridin (Ψ) umwandeln. Die H/ACA RNPs bestehen aus einer Leit-RNA und vier Proteinen, der katalytisch aktiven Pseudouridylase Cbf5, Nhp2, Gar1 und Nop10. Die Leit-RNA besteht in Eukaryoten konserviert aus zwei Haarnadelstrukturen, die von einem H-Box oder ACA-Box Sequenzmotiv gefolgt sind. In jeder dieser Haarnadeln befindet sich ein ungepaarter Bereich, die sogenannte Pseudouridylierungstasche, wo durch komplementäre Basenpaarung die Ziel-RNA gebunden wird. Fehlerhafte H/ACA RNPs können beim Menschen zu schweren Krankheiten wie verschiedenen Krebsarten oder dem Knochenmarksversagen Dyskeratosis congenita führen, aber sie bieten auch Möglichkeiten zum Einsatz als Therapiemethode. In dieser Arbeit wurde hauptsächlich der zweiteilige Aufbau der H/ACA RNPs untersucht.
Dafür wurden zunächst die einzelnen Komponenten hergestellt werden. Cbf5, Nop10 und Gar1 wurden zusammen heterolog in E. coli exprimiert und gereinigt. Außerdem wurden mehrere Deletionsvarianten von Gar1 hergestellt. Zusätzlich wurde die Leit-RNA unmarkiert über T7 Transkription synthetisiert, sowie sechs verschiedene FRET-Konstrukte mit verschiedenen Markierungschemas der Fluorophore Cy3 und Cy5 über DNA-geschiente Ligation. Anschließend wurde über Größenausschlusschromatographie und radioaktiven Aktivitätsassays geprüft, dass sich die aktiven H/ACA RNPs in vitro aus den einzelnen Komponenten rekonstituieren lassen.
In smFRET Experimenten wurden einzelne Haarnadelstrukturen mit dem zweiteiligen Komplexen verglichen. Dabei konnte gezeigt werden, dass die H3 Haarnadel durch die Anwesenheit von H5 dynamischer und heterogener wurde, während H5 überwiegend unbeeinflusst war. Außerdem konnte die dreidimensionale Orientierung der Haarnadelstrukturen in verschiedenen Assemblierungsschritten mittels smFRET untersucht werden. Hier deutete sich an, dass in Abwesenheit von Proteinen beide Haarnadeln eher entgegengesetzt stehen als in einer parallelen Konformation. Cbf5 scheint den Linker zwischen den Beiden auszustrecken bzw. zu orientieren und die Haarnadelstrukturen etwas gegeneinander zu neigen. Ein Zusammenspiel von Nhp2 und Gar1 war nötig um die oberen Bereiche der Haarnadeln zusammenzuziehen. Es konnte auch ein Modell für den vollen H/ACA RNP vorgeschlagen werden. Im kompletten Komplex könnte das Zusammenziehen der Haarnadelstrukturen durch Nhp2 und Gar1 mit dem Effekt von Cbf5 konkurrieren und könnte hauptsächlich den oberen Bereich von H3 betreffen. Zum Schluss wurde das Zusammenspiel von Gar1 und Nhp2 auf eine Abhängigkeit von den RGG Domänen von Gar1 hin untersucht. Hier besteht möglicherweise eine Hierarchie, die eine Kooperativität von den N- und C-terminalen Domänen benötigt.
Das zweite Projekt befasst sich mit dem Guanidin-II Riboschalter aus E. coli. Der Riboschalter kann das toxische Molekül Guanidinium (Gdm+) spezifisch in seiner Aptamerdomäne binden und dadurch die Genexpression von Proteinen zur Detoxifizierung von Gdm+ aktivieren. Der Riboschalter besteht aus zwei Haarnadelstrukturen, mit einer Schleife, die aus der Sequenz ACGR besteht, wobei R ein Purin ist. In einem vorgeschlagenen Modell soll die Ribosomenbindestelle (Shine-Dalgarno Sequenz) in Abwesenheit von Ligand mit dem Linker komplementär Basenpaaren und so die Translation verhindern. Mit Ligand würde sich dann eine Schleifen-Schleifen Interaktion mit den beiden CG Basen ausbilden, wodurch die Anti-Shine-Dalgarno Sequenz nicht mehr zugänglich wäre. Bisherige Studien arbeiteten zumeist nur mit der Aptamerdomäne, den einzelnen Haarnadeln oder noch kleineren Elementen. In dieser Arbeit wurden die Strukturdynamiken von verschiedenen Längen, auch mit der Expressionsplatform, untersucht. Außerdem wurden verschiedene Mutationen analysiert und die Effekte auf den Riboschalter in seiner natürlichen Umgebung in E. coli.
Zunächst mussten insgesamt 24 FRET-Konstrukte hergestellt werden, die sich in Länge, Markierungsschema und Mutationen unterschieden. Hierfür wurde DNA-geschiente Ligation verwendet. Dank der verschiedenen Fluorophorpositionen konnte ein konformationelles Modell für die Aptamerdomäne vorgeschlagen werden. In diesem Modell könnte in Abwesenheit von Ionen das Aptamer offen vorliegen. Durch Mg2+ würde sich bereits eine lockere Schleifen-Schleifen Interaktion ausbilden. Zusätzlich deuten die Ergebnisse auf eine neue Konformation hin, der stabilisierten Schleifen-Schleifen Interaktion, bei der der Linker zusätzlich mit den Haarnadelstrukturen interagiert, beispielswese mit den Purinen an der vierten Schleifenposition...
