Rapid alterations in protein expression are commonly regulated by adjusting translation. In addition to cap-dependent translation, which is e.g. induced by pro-proliferative signaling via the mammalian target of rapamycin (mTOR)-kinase, alternative modes of translation, such as internal ribosome entry site (IRES)-dependent translation, are often enhanced under stress conditions, even if cap-dependent translation is attenuated. Common stress stimuli comprise nutrient deprivation, hypoxia, but also inflammatory signals supplied by infiltrating immune cells. Yet, the impact of inflammatory microenvironments on translation in tumor cells still remains largely elusive. In the present study, we aimed at identifying translationally deregulated targets in tumor cells under inflammatory conditions. Using polysome profiling and microarray analysis, we identified cyp24a1 (1,25-dihydroxyvitamin D3 24-hydroxylase) to be translationally upregulated in breast tumor cells co-cultured with conditioned medium of activated monocyte-derived macrophages (CM). Using bicistronic reporter assays, we identified and validated an IRES within the 5′ untranslated region (5′UTR) of cyp24a1, which enhances translation of cyp24a1 upon CM treatment. Furthermore, IRES-dependent translation of cyp24a1 by CM was sensitive to phosphatidyl-inositol-3-kinase (PI3K) inhibition, while constitutive activation of Akt sufficed to induce its IRES activity. Our data provide evidence that cyp24a1 expression is translationally regulated via an IRES element, which is responsive to an inflammatory environment. Considering the negative feedback impact of cyp24a1 on the vitamin D responses, the identification of a novel, translational mechanism of cyp24a1 regulation might open new possibilities to overcome the current limitations of vitamin D as tumor therapeutic option.
The disintegrin and metalloproteinases ADAM10 and ADAM17 are regarded as the most important α-secretases involved in the physiological processing of amyloid precursor protein (APP) in brain. Since it has been suggested that processing of APP by α-secretases could be involved in the reorganization of the brain following injury, we studied mRNA expression of the two α-secretases Adam10 and Adam17, the ß-secretase Bace1, and the App-gene family (App, Aplp1, Aplp2) in the dentate gyrus of the mouse following entorhinal denervation. Using laser microdissection, tissue was harvested from the outer molecular layer and the granule cell layer of the denervated dentate gyrus. Expression levels of candidate genes were assessed using Affymetrix GeneChip Mouse Gene 1.0 ST arrays and reverse transcription-quantitative PCR, revealing an upregulation of Adam10 mRNA and Adam17 mRNA in the denervated outer molecular layer and an upregulation of Adam10 mRNA and App mRNA in the dentate granule cell layer. Immunolabeling for ADAM10 or ADAM17 in combination with markers for astro- and microglia revealed an increased labeling of ADAM10 and ADAM17 in the denervated outer molecular layer that was associated with reactive astrocytes but not with microglia. Collectively, these data show that denervation affects the expression level of APP and its two most important α-secretases. This suggests that APP-processing could be shifted towards the non-amyloidogenic pathway in denervated areas of the brain and, thus, towards the formation of neuroprotective APP cleavage products, such as APPsα.
Disruption of the renal endothelial integrity is pivotal for the development of a vascular leak, tissue edema and consequently acute kidney injury. Kidney ischemia amplifies endothelial activation and up-regulation of pro-inflammatory mechanisms. After restoring a sufficient blood flow, the kidney is damaged through complex pathomechanisms that are classically referred to as ischemia and reperfusion injury, where the disruption of the inter-endothelial connections seems to be a crucial step in this pathomechanism. Focusing on the molecular cell-cell interaction, the fibrinopeptide Bβ15–42 prevents vascular leakage by stabilizing these inter-endothelial junctions. The peptide associates with vascular endothelial-cadherin, thus preventing early kidney dysfunction by preserving blood perfusion efficacy, edema formation and thus organ dysfunction. We intended to demonstrate the early therapeutic benefit of intravenously administered Bβ15–42 in a mouse model of renal ischemia and reperfusion. After 30 minutes of ischemia, the fibrinopeptide Bβ15–42 was administered intravenously before reperfusion was commenced for 1 and 3 hours. We show that Bβ15–42 alleviates early functional and morphological kidney damage as soon as 1 h and 3 h after ischemia and reperfusion. Mice treated with Bβ15–42 displayed a significantly reduced loss of VE-cadherin, indicating a conserved endothelial barrier leading to less neutrophil infiltration which in turn resulted in significantly reduced structural renal damage. The significant reduction in tissue and serum neutrophil gelatinase-associated lipocalin levels reinforced our findings. Moreover, renal perfusion analysis by color duplex sonography revealed that Bβ15–42 treatment preserved resistive indices and even improved blood velocity. Our data demonstrate the efficacy of early therapeutic intervention using the fibrinopeptide Bβ15–42 in the treatment of acute kidney injury resulting from ischemia and reperfusion. In this context Bβ15–42 may act as a potent renoprotective agent by preserving the endothelial and vascular integrity.
Evidence from anatomical and functional imaging studies have highlighted major modifications of cortical circuits during adolescence. These include reductions of gray matter (GM), increases in the myelination of cortico-cortical connections and changes in the architecture of large-scale cortical networks. It is currently unclear, however, how the ongoing developmental processes impact upon the folding of the cerebral cortex and how changes in gyrification relate to maturation of GM/WM-volume, thickness and surface area. In the current study, we acquired high-resolution (3 Tesla) magnetic resonance imaging (MRI) data from 79 healthy subjects (34 males and 45 females) between the ages of 12 and 23 years and performed whole brain analysis of cortical folding patterns with the gyrification index (GI). In addition to GI-values, we obtained estimates of cortical thickness, surface area, GM and white matter (WM) volume which permitted correlations with changes in gyrification. Our data show pronounced and widespread reductions in GI-values during adolescence in several cortical regions which include precentral, temporal and frontal areas. Decreases in gyrification overlap only partially with changes in the thickness, volume and surface of GM and were characterized overall by a linear developmental trajectory. Our data suggest that the observed reductions in GI-values represent an additional, important modification of the cerebral cortex during late brain maturation which may be related to cognitive development.
Survival according to BRAF-V600 tumor mutations : an analysis of 437 patients with primary melanoma

The prognostic impact of BRAF-V600 tumor mutations in stage I/II melanoma patients has not yet been analyzed in detail. We investigated primary tumors of 437 patients diagnosed between 1989 and 2006 by Sanger sequencing. Mutations were detected in 38.7% of patients and were associated with age, histological subtype as well as mitotic rate. The mutational rate was 36.7% in patients with disease-free course and 51.7% in those with subsequent distant metastasis (p = 0.031). No difference in overall survival (p = 0.119) but a trend for worse distant-metastasis-free survival (p = 0.061) was observed in BRAF mutant compared to BRAF wild-type patients. Independent prognostic factors for overall survival were tumor thickness, mitotic rate and ulceration. An interesting significant prognostic impact was observed in patients with tumor thickness of 1 mm or less, with the mutation present in 6 of 7 patients dying from melanoma. In conclusion, no significant survival differences were found according to BRAF-V600 tumor mutations in patients with primary melanoma but an increasing impact of the mutational status was observed in the subgroup of patients with tumor thickness of 1 mm or less. A potential role of the mutational status as a prognostic factor especially in this subgroup needs to be investigated in larger studies.
We consider versions of the FIND algorithm where the pivot element used is the median of a subset chosen uniformly at random from the data. For the median selection we assume that subsamples of size asymptotic to c⋅nα are chosen, where 0<α≤12, c>0 and n is the size of the data set to be split. We consider the complexity of FIND as a process in the rank to be selected and measured by the number of key comparisons required. After normalization we show weak convergence of the complexity to a centered Gaussian process as n→∞, which depends on α. The proof relies on a contraction argument for probability distributions on càdlàg functions. We also identify the covariance function of the Gaussian limit process and discuss path and tail properties.
Natural products (NPs) have been a rich source for pharmaceutically used anti-infectives and other drugs. However, the application of anti-infectives inevitably causes the development of resistant and multiresistant pathogens, which have to be treated with novel anti-infectives. The industrial research for novel anti-infectives has been concentrating on members of the bacterial Actinomycetales for a long time. Due to several reasons, e.g. the rediscovery of already known NPs, pharmaceutical companies abandoned their NP-research and focused on drug development based on combinatorial chemistry. However, the limited structural diversity of merely synthetic compound libraries has not been a fruitful source for bioactive compounds. Hence the discovery of novel bioactive NPs as a source for anti-infectives is still of economical and humanitarian interest and will remain to be an important branch of research in the future. One strategy to circumvent the rediscovery of bioactive NPs is the analysis of yet unexplored bacterial taxa. Based on this assumption, this work aimed at the discovery of novel NPs from the entomopathogenic bacterial genera Xenorhabdus and Photorhabdus and other promising taxa, as well as the investigation of their biosynthesis. ...
The article introduces a research project financed by the Academy of Sciences and Literature Mainz began in 2013 and will extend over an 18-year period. It aims at producing a historical-semantic dictionary elucidating central terms of the School of Salamanca's discourses and their significance for modern political theory and jurisprudence. The project's fundament will be a digital corpus of important texts from the School of Salamanca which will be linked up with the dictionary's online version. By making the source corpus accessible in searchable full text (as well as in high quality digital images), the project is creating a new research tool with exciting possibilities for further investigations. The dictionary will be a valuable source of information for the interdisciplinary research carried out in this field.
Experimente und Simulationen zur Filterung und Injektion in einen toroidalen Ionenspeicherring

Die in dieser Arbeit durchgeführten theoretischen Untersuchungen, Simulationen und Experimente hatten das Ziel, die Erforschung der Injektion in sowie des Transports von Ionenstrahlen bestehend aus verschiedenen, einzelnen Wasserstoffionenspezies durch toroidale Magnetfeldstrukturen zu ermöglichen. Dazu wurde im Rahmen dieser Arbeit ein neues Filter- und Detektorsystem für Wasserstoffionenstrahlen – ein Filterkanal – entwickelt, aufgebaut und untersucht. Zunächst wurde mittels theoretischer Betrachtungen das Konzept des Kanals und die Funktionalität der Filterung sowie Vermessung der Strahlzusammensetzung analysiert. Danach wurde der Kanal mit Hilfe von Simulationen ausgelegt und anschließend nach genauen Vorgaben gebaut. Gleichzeitig wurde mittels weiterer Simulationen das Strahlverhalten im Kanal genauer analysiert und zusätzlich die Auswirkungen verschiedener Parameter auf das Filterverhalten des Kanals untersucht. Anschließend wurde der zusammengebaute Kanal mittels Experimenten erforscht. Dabei wurden mehrere Experimente mit verschiedenen Quellen- und Strahlparametern durchgeführt, wobei vor allem die Filtereigenschaften sowie die Vermessung der Strahlzusammensetzung umfangreich untersucht und im Bezug auf die Funktionalität des Filterkanals diskutiert wurden.
Des Weiteren befasste sich diese Arbeit mit dem Aufbau des Injektionsexperiments zur Untersuchung der Injektion eines Wasserstoffionenstrahls zwischen zwei Toroidsegmente. Dazu wurde ein höhenverstellbarer Injektor samt Hochspannungsterminal und Peripherie entwickelt, aufgebaut und in Betrieb genommen. Dieser besteht aus einer Volumenionenquelle, einer neu entwickelten Faraday-Tasse sowie einem Solenoid. Die Faraday-Tasse, welche zur Vermessung des Quellenstroms im neuen Injektor sowie zusätzlich zur Vermessung des Ionenstrahlstroms hinter dem Filterkanal eingesetzt wird, wurde dabei im Rahmen dieser Arbeit entwickelt, experimentell durchgemessen und auf ihre Funktionalität geprüft. Im Zusammenhang mit dem Hochspannungsterminal für den neuen Injektor wurden die verschiedenen Möglichkeiten für die Verschaltung einer Volumenionenquelle mit dem dazugehörigen Hochspannungsterminal diskutiert und der Zusammenhang zur Strahlenergie hergestellt.
Mittels theoretischer Rechnungen wurde der aufgrund der RxB Drift benötigte Höhenunterschied zwischen Injektionskanal und Transportkanal für das Injektionsexperiment bestimmt und der neue Injektor auf den errechneten Bereich vorkonfiguriert. Zusätzlich wurde mittels Simulationen die optimale Strahlenergie bestimmt, bei der der Strahl beim Transport durch den Transportkanal angepasst ist und somit die Auswirkung der Driftsektion zwischen den beiden Toroidsegmenten sowie die Beeinflussung durch das Injektionssystem am geringsten sein sollten. In diesem Zusammenhang wurde auch auf die möglichen Geometrien zum Aufbau des Injektionsexperiments eingegangen und mittels der angesprochenen Untersuchungen eine Geometrie für den Aufbau des Injektionssystems festgelegt.
Die hier vorgelegte empirische Untersuchung der Fokuspartikeln im Georgischen zeichnet sich u.a. durch die sprach¬immanente Tatsache aus, dass die Fokusstrukturen im Georgischen mit expliziten Partikeln markiert werden können. Die in dieser Arbeit untersuchten Fokuspartikelgruppen ( ġa, c und c+ḳi) sind entsprechend den semantischen Implikationen der Restriktion, Addition und der Skalierung gegliedert worden.
Trotz gewisser Unterschiede im Einzelnen ergab sich folgendes gemeinsames Modell für die Stellungseinschränkungen in Relation zum Prädikatsverb:
• Durch Fokuspartikeln fokussierte Wörter stehen im Georgischen in der Regel unmittelbar vor dem Prädikatsverb.
• Die Skopi der Fokuspartikeln (wenn die fokusmarkierten Worte grammatische Köpfe der NPs sind) stehen im Georgischen in der Regel vor dem Prädikatsverb.
• Die nächstmögliche optimale Interpretationsposition für fokusmarkierte Wörter ist in der Regel die unmittelbare Verbnachstellung.
• Die nächstmögliche optimale Interpretationsposition der
Fokusgruppe ist in der Regel die unmittelbare Verbnachstellung.
Aufgrund der herausgearbeiteten Stellungseinschränkungen entwerfe ich das pragmatische Modell der informationsgliedernden Verbfinalität als Basisabfolge im georgischen Satz.
In der Literatur hat sich die Bestimmung der Erzählperspektive längst etabliert, aber genauso manifestiert sich auch in den visuellen Medien ein Erzähler, der das Geschehen zwar weniger mittels der Sprache, dafür im Wesentlichen über Bilder präsentiert. Durch die jeweils darin enthaltene Darstellung und die Verknüpfung der einzelnen Bilder sind diese Ausdruck einer bewussten Formung der Handlung. Der Begriff des Erzählens beschränkt sich somit nicht nur auf die Literatur, sondern trifft auch auf Visualisierungen jeglicher Art zu, da in diesen Fällen ebenso eine Instanz identifiziert werden kann, die hinter der Auswahl der Handlungselemente steht und diese perspektivisch formt. Dennoch kann der Erzähler hier nicht mehr, wie in der Literatur, im Sinne eines "Sprechers" aufgefasst werden, sondern versteht sich eher als abstraktes Konzept. Insbesondere in intermedialer Hinsicht stellt die Übertragung der Erzählperspektive von einem literarischen Text auf ein visuelles Medium eine Herausforderung dar, denn aufgrund des Medienwechsels verändern sich die Bedingungen des Erzählens. Umso schwieriger ist die Übertragung, wenn der literarische Text keinen durchweg logischen Handlungsverlauf aufweist, sondern Widersprüche und Uneindeutigkeiten im Erzählvorgang und in der Erzählperspektive beinhaltet, wie es auf Franz Kafkas 1914 entstandene Erzählung "In der Strafkolonie" zutrifft. Dieser Text dient zudem, wie auch viele andere von Kafkas Werken, als Projektionsfläche diverser, nicht selten konträrer, Deutungsansätze, die sich in der Forschungsliteratur herausgebildet haben. So wurden zahlreiche religiöse Bezüge in der Erzählung entdeckt, genauso wurde der Text aber als realistisch und somit als Kritik an Kolonialismus und Krieg verstanden. Bezug nehmend auf Kafkas Biographie und das Thema Strafe, das in zahlreichen seiner Werke zum Ausdruck kommt, wurde die Erzählung oftmals in psychoanalytischer Hinsicht gedeutet. Andererseits wurde die beschriebene Handlung auch weniger ernst genommen und vor allem auf die grotesken und ironischen Elemente verwiesen. Im Hinblick auf die unterschiedlichen Interpretationsansätze sind auch die Möglichkeiten der visuellen Umsetzung entsprechend vielfältig.
In dieser Arbeit wird anhand von drei Kurzfilmen und einem Comic, unter Berücksichtigung der spezifischen medialen Bedingungen, die Übertragung der Erzählung ins Visuelle analysiert. Dabei soll gezeigt werden, inwiefern die jeweilige Erzählperspektive die visuelle Wahrnehmung in den Filmen und im Comic bestimmt. In diesem Zusammenhang wird auch zu sehen sein, welche Deutung der Erzählung sich in der Visualisierung manifestiert und durch den Erzähler besonders hervorgehoben wird. Nicht zuletzt stellt sich dabei auch die Frage nach der Art der Umsetzung, d.h. ob bei der Adaption die Textnähe und der Inhalt im Vordergrund stehen oder ob vielmehr die Intention verfolgt wird, mit der Visualisierung der Erzählung ein neues und eigenes künstlerisches Werk zu schaffen, das dem Originaltext auf einer tiefergehenden Ebene begegnet.
Eryphanis zolvizora (Hewitson, 1877) is a rare Andean endemic butterfly, described from Bolivia, which has been historically classified either as a unique species, or as part of a group of three allopatric species from Bolivia, Ecuador and Colombia. In this paper, the group is revised using more than 200 specimens housed in 35 European and North and South American public and private collections. For the first time, the presence of the group in Western Ecuador and Venezuela is confirmed, and important data on Peruvian populations are provided. In some populations, individual variations of genitalia are observed. Nevertheless, male genitalia allow the distinction of four geographical groups. Considering also habitus characters, eight taxa are distinguished and considered to be subspecies, of which five are new: Eryphanis zolvizora inca ssp. nov., Eryphanis zolvizora chachapoya ssp. nov., Eryphanis zolvizora casagrande ssp. nov., Eryphanis zolvizora reyi ssp. nov., and Eryphanis zolvizora isabelae ssp. nov. In the present state of knowledge, these taxa are allopatric, except for a possible geographic overlap in central Peru, where data are insufficient to prove sympatry. The “several subspecies vs. several species” dilemma is discussed, considering its impact for conservation action and policies.
This is the first English edition of Tamara Mcheidze's monograph on the spiders of Georgia (Mcheidze 1997; in alternative transliteration: Mkheidze 1997), containing taxonomic, faunistic and ecological information on 362 spider species recorded by T. Mcheidze in Georgia between the 1920s and 1992. The English edition resembles the original text, with a preface, taxonomic comments and a part with supplementary information on the locations, a location map and an index provided by the editor.
The administration of intravenous fluid to critically ill patients is one of the most common but also one of the most fiercely debated interventions in intensive care medicine. During the past decade, a number of important studies have been published which provide clinicians with improved knowledge regarding the timing, the type and the amount of fluid they should give to their critically ill patients. However, despite the fact that many thousands of patients have been enrolled in these trials of alternative fluid strategies, consensus remains elusive and practice is widely variable. Early adequate resuscitation of patients in shock followed by a restrictive strategy may be associated with better outcomes. Colloids such as modern hydroxyethyl starch are more effective than crystalloids in early resuscitation of patients in shock, and are safe when administered during surgery. However, these colloids may not be beneficial later in the course of intensive care treatment and should best be avoided in intensive care patients who have a high risk of developing acute kidney injury. Albumin has no clear benefit over saline and is associated with increased mortality in neurotrauma patients. Balanced fluids reduce the risk of hyperchloraemic acidosis and possibly kidney injury. The use of hypertonic fluids in patients with sepsis and acute lung injury warrants further investigation and should be considered experimental at this stage. Fluid therapy impacts relevant patient-related outcomes. Clinicians should adopt an individualized strategy based on the clinical scenario and best available evidence. One size does not fit all.
Four taxa belonging to the complex of species around Achnanthidium minutissimum were found during the ongoing taxonomic revision of the Antarctic freshwater and limno-terrestrial diatom flora. Two taxa were previously described as Achnanthidium lailae and A. sieminskae. Two others were formerly identified as A. minutissimum but detailed light and scanning electron microscopical observations revealed sufficient morphological differences compared to the type of A. minutissimum, to justify their separation and description as new taxa: Achnanthidium indistinctum and A. maritimo-antarcticum. The morphology and ecology of all four taxa are discussed comparing the species with morphologically similar taxa. The biogeographical consequences of the splitting of the former A. minutissimum complex in the Antarctic Region are discussed.
In this paper, we propose a novel approach on how to estimate systemic risk and identify its key determinants. For all US financial companies with publicly traded equity options, we extract their option-implied value-at-risks (VaRs) and measure the spillover effects between individual company VaRs and the option-implied VaR of an US financial index. First, we study the spillover effect of increasing company risks on the financial sector. Second, we analyze which companies are most affected if the tail risk of the financial sector increases. We find that key accounting and market valuation metrics such as size, leverage, balance sheet composition, market-to-book ratio and earnings have a significant influence on the systemic risk profile of a financial institution. In contrast to earlier studies, the employed panel vector autoregression (PVAR) estimator allows for a causal interpretation of the results.
Die Texte der vorliegenden Arbeit sind Stationen auf dem Weg zu einer normativen Theorie der romantischen Liebe. Was hier unter einer normativen Theorie der romantischen Liebe verstanden wird, gründet auf folgenden Annahmen über das Liebesphänomen und lässt sich in Bezug auf sie kurz zusammenfassen: das Phänomen der romantischen Liebe – sowie das Liebesphänomen als Ganzes – bildet ein Kontinuum. An dem einen Ende dieses Spektrums gibt es gute, an dem anderen Ende schlechte Ausprägungen der Liebe. Die guten Ausprägungen der Liebe und die Liebesbeziehungen, die daraus erwachsen, sind für die liebenden und für die geliebten Personen ein zentraler Bestandteil eines guten und glücklichen Lebens. Die schlechten Ausprägungen der Liebe und die aus ihnen hervorgehenden Liebesbeziehungen untergraben umgekehrt für die liebenden und für die geliebten Personen die Möglichkeit eines guten und glücklichen Lebens. Eine normative Theorie der Liebe, wie sie in dieser Arbeit aus verschiedenen Blickwinkeln formuliert wird, versucht sowohl die guten als auch die schlechten Ausformungen der Liebe zu identifizieren, sie in ihrer Phänomenologie darzustellen, ihren positiven oder negativen Charakter zu bestimmen und ihren negativen oder positiven Beitrag zum guten Leben auszubuchstabieren.
Einen ersten Schritt in die skizzierte theoretische Richtung wird in dem Rahmen einer Lektüre von Platons Phaidros unternommen, die den ersten Teil der Arbeit bildet. Die erste Hälfte des Phaidros ist dem Phänomen der erotischen Liebe gewidmet, sowie es innerhalb einer zu Platons Zeiten anerkannten Beziehungsform, nämlich der „Knabenliebe“, praktiziert wurde. In den drei Liebesreden, die in der ersten Hälfte des Dialogs vorgetragen werden, werden zuerst eine schlechte und am Ende eine gute und „göttliche“ Ausprägung des Eros aus jeweils verschiedenen normativen Perspektiven dargestellt. In Sokrates Kritik des schlechten Eros zeigt sich dieser nämlich als eine Spezies der hybris („Frevel“), die im Gegensatz zu der Disposition der Seelenkräfte steht – die Besonnenheit – die zum „Besten“, d.h. zur Tugend führt. In der Lobrede an den Eros wird die Liebe hingegen in dem Rahmen eines Mythos als ein zentraler Aspekt eines guten und glücklichen Lebens mit allen möglichen poetischen und philosophischen Mitteln dargestellt und gelobt. Der gute Eros wird als eine Umwälzung und Transformation der ganzen Seele, als die Wiederherstellung ihrer verlorenen Einheit durch die Schönheit des Geliebten charakterisiert. Beide Liebende vermögen im Rahmen einer lebenslangen Freundschaft, durch eine Einsicht und Erkenntnis des jeweils anderen, durch eine beidseitige Prägung und „Ausbildung“, die besseren Teile der Seele zur Entfaltung zu bringen.
Der zweite Teil der Arbeit beginnt mit einer literarischen Perspektive auf die romantische Liebe. Der Ausgangspunkt dabei ist eine Darstellung von Stendhals Theorie der romantischen Liebe, die „Kristallisationstheorie“ der Liebe, die der Autor in seinem Über die Liebe formuliert hat. Eine zentrale These der Kristallisationstheorie der Liebe lautet, dass die geliebte Person weitestgehend ein Trugbild, ein Phantasiekonstrukt des Liebenden ist. Liebende sehen ihre Geliebten nicht so, wie sie sind, sondern wie sie sich nach ihren Wünschen zurechtmachen. In diesem Rahmen wird der Versuch unternommen, die zentrale These der Kristallisationstheorie im Zusammenhang mit Formen des Selbstbetrugs zu erläutern und sie plausibel zu machen. Zu diesem Zweck werden philosophische Beiträge herangezogen, die das richtige Verständnis vom Phänomen des Selbstbetrugs herauszuarbeiten versuchen. Dabei werden mögliche psychische Mechanismen des Selbstbetrugs identifiziert und untersucht, die insgesamt bewirken, dass die Liebenden die geliebte Person nicht so sehen, wie sie ist, sondern sie stattdessen durch die Zerrbrille ihrer Wünsche und Begierden wahrnehmen.
Nachdem im dritten Teil der Arbeit Platons und Stendhals Liebestheorien auf ihre Unterschiede hin untersucht werden, ist im vierten und längsten Teil Gegenstand der Arbeit, eine normative Theorie der romantischen Liebe entlang drei Grunddimensionen der personalen Liebe in Grundrissen zu skizzieren: das Liebessubjekt (oder die Liebenden), das Liebesobjekt (oder die Geliebten) und die Liebesbeziehung (oder das Liebespaar). Diese Aufgabe wird in einer Auseinandersetzung mit neueren Beiträgen der Philosophie der Liebe sowie sie von solchen Autoren wie Harry Frankfurt und David Velleman artikuliert wird, aber auch im Rückgriff auf einige Grundideen aus Platons Phaidros sowie im Zusammenhang mit literarischen Werken wie Prousts Á la recherche oder Flauberts Madame Bovary und mit Filmen (Michael Hanekes Liebe und Derek Cianfrances Blue Valentine) in Angriff genommen. Dabei liegt der Akzent auf einer Erläuterung grundlegender Aspekte der Liebenden oder des Liebessubjekts: der Wille, die Gefühle, der Blick bekommen in der Liebe eine besondere Prägung, spielen miteinander zusammen und bilden zentrale Elemente der liebenden Einstellung von Personen. Eine in Platon inspirierte Grundidee dabei lautet, dass Liebe sämtliche geistige Kräfte einbezieht, sodass die Begrenzung auf nur eins dieser Elemente, so wie sie durch viele der behandelten Autoren vorgenommen wird, strikt abzulehnen ist. Stattdessen wird ein integrativer Ansatz vorgeschlagen und verfolgt. Die Schilderung dieser Aspekte der Liebe folgt dem bereits erwähnten platonischen Gedanken, dass es sowohl gute als auch schlechte Ausprägungen der Liebe gibt, sodass immer wieder gute und schlechte Formen der jeweils thematisierten Aspekte der Liebe unterschieden werden.
