W±-boson production in p–Pb collisions at √sNN = 8.16 TeV and Pb–Pb collisions at √sNN = 5.02 TeV

The production of the W± bosons measured in p–Pb collisions at a centreof-mass energy per nucleon–nucleon collision √sNN = 8.16 TeV and Pb–Pb collisions at √sNN = 5.02 TeV with ALICE at the LHC is presented. The W± bosons are measured via their muonic decay channel, with the muon reconstructed in the pseudorapidity region −4 < ηµ lab < −2.5 with transverse momentum p µ T > 10 GeV/c. While in Pb–Pb collisions the measurements are performed in the forward (2.5 < yµ cms < 4) rapidity region, in p–Pb collisions, where the centre-of-mass frame is boosted with respect to the laboratory frame, the measurements are performed in the backward (−4.46 < yµ cms < −2.96) and forward (2.03 < yµ cms < 3.53) rapidity regions. The W− and W+ production cross sections, leptoncharge asymmetry, and nuclear modification factors are evaluated as a function of the muon rapidity. In order to study the production as a function of the p–Pb collision centrality, the production cross sections of the W− and W+ bosons are combined and normalised to the average number of binary nucleon–nucleon collision hNcolli. In Pb–Pb collisions, the same measurements are presented as a function of the collision centrality. Study of the binary scaling of the W±-boson cross sections in p–Pb and Pb–Pb collisions is also reported. The results are compared with perturbative QCD calculations, with and without nuclear modifications of the Parton Distribution Functions (PDFs), as well as with available data at the LHC. Significant deviations from the theory expectations are found in the two collision systems, indicating that the measurements can provide additional constraints for the determination of nuclear PDFs and in particular of the light-quark distributions.
Bislang sind die strukturellen Voraussetzungen für die Selektivität von Agonisten an den Retinoid Rezeptor Subtypen RXRα, RXRβ und RXRγ kaum erforscht, obwohl RXR-Modulatoren, die eine Subtypen-Präferenz aufweisen, aufgrund der unterschiedlichen Expressionsmuster der Subtypen Gewebe-spezifische Effekte vermitteln und somit Nebenwirkungen verringern könnten. Der Grund dieser Forschungslücke liegt teilweise darin, dass die Entwicklung Subtypen-selektiver RXR-Agonisten aufgrund der enormen strukturellen Ähnlichkeit der Ligandbindestellen in den RXR-Subtypen - alle Aminosäuren, die die Bindungsstellen bilden sind identisch - als unerreichbar angesehen wurde. Die Entdeckung des Naturstoffs Valerensäure als RXR-Agonist mit ausgeprägter Präferenz für den RXRβ-Subtyp hat jedoch gezeigt, dass Subtypen-selektive RXR-Modulation möglich ist249 und SAR-Studien an unterschiedlichen RXR-Ligand-Chemotypen haben in der Folge bestätigt, dass die Entwicklung von RXR-Liganden mit Subtypen-Präferenz erreicht werden kann.
Auf der Basis von Valerensäure und der in früheren Arbeiten entwickelten RXR-Agonisten wurden in dieser Arbeit Strukturmodifikationen identifiziert, die zu einer RXR-Subtypen-Präferenz beitragen. Durch die Verschmelzung dieser Strukturelemente ist es gelungen, einen neuen RXR-Agonist-Chemotyp (A) zu entwerfen, der durch strategische Methylierung und weitere Strukturmodifikationen zur Präferenz für jeden Subtyp optimiert werden konnte.
In einem Adipozyten-Differenzierungsexperiment konnte gezeigt werden, dass RXRα der wichtigste Heterodimer-Partner von PPARγ während der Adipogenese ist. Ferner unterstrich diese biologische Untersuchung das Potenzial von 99, 103 und 105 als Subtyp-präferentielle RXR-Agonisten in vitro Experimenten zu dienen.
Auf der Grundlage dieser Ergebnisse wurde eine mögliche Rolle von Acrylsäurepartialstrukturen natürlicher RXR-Liganden basierend auf dem zuvor entwickelten Chemotyp untersucht. Hierzu wurden das α-Methylacrylsäuremotiv des Naturstoffs Valerensäure (18) und das β-Methylacrylsäuremotiv des endogenen RXR-Agonisten 9-cis-Retinsäure in den Chemotyp A integriert (Chemotyp B), um die Rolle dieser Acrylsäuregruppen bei der Vermittlung der RXR-Subtypen-Selektivität zu untersuchen. Die Strukturmodifikationen an B zeigten, dass nur die α-Methyl-substituierte Acrylsäurekette toleriert bzw. von RXRβ präferiert wurde, was die RXR-Präferenz der Valerensäure (18) unterstützte.
In dieser Arbeit konnte gezeigt werden, dass RXR-Liganden mit Subtypen-Präferenz realisierbar sind und durch gezielte Strukturmodifikationen in ihrer Präferenz gesteuert werden können. Die Erkenntnisse zu den Struktur-Wirkungs-Beziehungen der neuen RXR-Agonist-Chemotypen A und B erweitern den Wissenstand über die strukturellen Voraussetzungen von RXR-Liganden für die Subtypen-Präferenz deutlich.
So abenteuerlich die Wege von deutschen Exilschriftstellerinnen und -schriftstellern des letzten Jahrhunderts waren, so verworren sind meist auch die Wege ihrer Nachlässe. Selten finden sich alle Manuskripte, Briefe und persönlichen Gegenstände an einem Ort versammelt. Häufig verteilen sich Nachlässe auf verschiedene Orte und Länder. Im schlimmsten Fall hat überhaupt niemand etwas aufbewahrt. Der Nachlass des Schriftstellers und Philosophen Hermann Borchardt (1888–1951) findet sich an zwei Standorten: im Deutschen Exilarchiv 1933–1945 in Frankfurt am Main, wohin ihn der verdienstvolle Exilforscher John M. Spalek überführte, und in der Rubenstein Rare Book & Manuscript Library in Durham, North Carolina. Ein unerwarteter Fund, den mein Kollege Christoph Hesse und ich dort machten, veranlasste uns, Borchardt mit einer Werkedition als wichtigen Schriftsteller des Exils zu würdigen.
Im 1. Teil werden zunächst religionsphilosophische, theologische und kulturtheoretische Voraussetzungen der Christian Songs behandelt.
Dazu werden die Begriffe Religion, Religionskultur, Kultur, Mythologie und Glaubensbilder geklärt. Im nächsten Schritt wird die protestantische Existenzper-spektive Martin Luthers ausführlicher dargestellt. Die Ausführlichkeit ergibt sich daraus, daß Martin Luthers Religionslehre das theologische Fundament der Chris-tians Songs ausmacht, sie aber zumeist in der vom Zeitgeist bestimmten Öffent-lichkeit nicht sonderlich bekannt ist oder neuerdings verzerrt dargestellt wird.
Abschließend wird, um den weltweiten Erfolg der Sangesbewegung zu verstehen, kurz deren Performance angesprochen.
Im 2. Teil werden ausgewählte Christian Songs insbesondere daraufhin unter-sucht und kommentiert, inwieweit sie das protestantische Existenzverständnis im Sinne Martin Luthers, insbesondere die theologischen Grundsätze sola gratia - allein aus Gnaden und sola fide – allein aus Glauben widerspiegeln.
Nitrogen pollution is a major constituent of global change, threatening local biodiversity, ecosystem services, and causing serious environmental damage. Specifically, in areas with heavy agricultural soil-use, excessive use of nitrogen fertilizer pollutes the groundwaters with nitrates, but also with ammonia and nitrites. Freshwater fish and other aquatic fauna are especially vulnerable to nitrites, which can cause massive mortalities at even low concentrations < 0.1 mg/l NO2- - N. Adaptation of fish to environments with relatively high concentrations of chemicals has occurred throughout the history of life, although contemporary evolution acts at a much more rapid pace. The growing use of land for mass agriculture and livestock industries in the last 50 years in the US has dramatically increased the nutrient loading into the surface and groundwaters. Nitrite poses a serious threat for freshwater fauna as it is rapidly up taken and disturbs ion homeostasis and blood gas transport in fish. In this study, we evaluated, by means of a laboratory experiment, the tolerance of fish to nitrite using six different populations of wild eastern mosquitofish (Gambusia holbrooki) from two regions, North FL and NC, and with different background nitrogen pollution histories. Mosquitofish females were exposed to nitrite in the lab, to either < 0.005 mg/l NO2- (control) or 0.3 mg/l NO2- for ten days and we assessed at the end of the exposure period their blood O2 transport capacity by measuring the concentration of four different types of hemoglobin, their total hematocrit, and their respiratory rates. Preliminary results show significant varying patterns in the response of the exposed fish, depending on the population source, as evidenced by their respiratory rates and the blood erythrocyte counts. Mortality was very low, and hemoglobin profiles indicate high tolerance of G. holbrooki to nitrite contamination – a factor supporting their invasion success in agriculturally dominated regions around the world
This cumulative dissertation contains four self-contained chapters on stochastic games and learning in intertemporal choice.
Chapter 1 presents an experiment on value learning in a setting where actions have both immediate and delayed consequences. Subjects make a series of choices between abstract options, with values that have to be learned by sampling. Each option is associated with two payoff components: One is revealed immediately after the choice, the other with one round delay. Objectively, both payoff components are equally important, but most subjects systematically underreact to the delayed consequences. The resulting behavior appears impatient or myopic. However, there is no inherent reason to discount: All rewards are paid simultaneously, after the experiment. Elicited beliefs on the value of options are in accordance with choice behavior. These results demonstrate that revealed impatience may arise from frictions in learning, and that discounting does not necessarily reflect deep time preferences. In a treatment variation, subjects first learn passively from the evidence generated by others, before then making a series of own choices. Here, the underweighting of delayed consequences is attenuated, in particular for the earliest own decisions. Active decision making thus seems to play an important role in the emergence of the observed bias.
Chapter 2 introduces and proves existence of Markov quantal response equilibrium (QRE), an application of QRE to finite discounted stochastic games. We then study a specific case, logit Markov QRE, which arises when players react to total discounted payoffs using the logit choice rule with precision parameter λ. We show that the set of logit Markov QRE always contains a smooth path that leads from the unique QRE at λ = 0 to a stationary equilibrium of the game as λ goes to infinity. Following this path allows to solve arbitrary finite discounted stochastic games numerically; an implementation of this algorithm is publicly available as part of the package sgamesolver. We further show that all logit Markov QRE are ε-equilibria, with a bound for ε that is independent of the payoff function of the game and decreases hyperbolically in λ. Finally, we establish a link to reinforcement learning, by characterizing logit Markov QRE as the stationary points of a game dynamic that arises when all players follow the well-established reinforcement learning algorithm expected SARSA.
Chapter 3 introduces the logarithmic stochastic tracing procedure, a homotopy method to compute stationary equilibria for finite and discounted stochastic games. We build on the linear stochastic tracing procedure (Herings and Peeters 2004), but introduce logarithmic penalty terms as a regularization device, which brings two major improvements. First, the scope of the method is extended: it now has a convergence guarantee for all games of this class, rather than just generic ones. Second, by ensuring a smooth and interior solution path, computational performance is increased significantly. A ready-to-use implementation is publicly available. As demonstrated here, its speed compares quite favorable to other available algorithms, and it allows to solve games of considerable size in reasonable times. Because the method involves the gradual transformation of a prior into equilibrium strategies, it is possible to search the prior space and uncover potentially multiple equilibria and their respective basins of attraction. This also connects the method to established theory of equilibrium selection.
Chapter 4 introduces sgamesolver, a python package that uses the homotopy method to compute stationary equilibria of finite discounted stochastic games. A short user guide is complemented with discussion of the homotopy method, the two implemented homotopy functions logit Markov QRE and logarithmic tracing, and the predictor-corrector procedure and its implementation in sgamesolver. Basic and advanced use cases are demonstrated using several example games. Finally, we discuss the topic of symmetries in stochastic games.
This cumulative dissertation examines learning in chemistry laboratories, focusing on the challenges and benefits of problem-based learning (PBL) for novices in the lab. It addresses the lack of consistent understanding about what should be learned in labs and why it's important. The research aims to understand what students learn, how they learn, and how lab learning can be improved.
A central concept in PBL labs is Information Literacy, defined as a sociocultural practice enabling learners to identify and use information sources within a specific context as legitimized by the practice community.
The first publication, Wellhöfer and Lühken (2022a), investigates the relationship between PBL and learner motivation. It identifies factors that can foster students' intrinsic motivation in a PBL lab. Autonomy is found to be a key factor, increasing student motivation and presenting a model of the autonomous scientific process. This model involves four steps: information acquisition, designing and applying experimental procedures, experimental feedback, and autonomous process optimization. The results suggest that intrinsic motivation in PBL labs can be enhanced by enabling students to independently execute these steps.
The second publication, Wellhöfer and Lühken (2022b), examines the information process students undergo during their first PBL lab. Using a sociocultural framework, it explores Information Literacy to understand students' handling of information and their perceptions of the information process. The findings reveal that in PBL labs, developing a practical, applicable experimental procedure is crucial for problem-solving and significantly shapes the information-acquisition process. This process is iterative, influenced by new information, leading to more precise information needs. Students assess information quality based on its usefulness for their problem, implementability (considering cognitive understanding, available equipment, and psychomotor skills), and safety.
Furthermore, the role of privileged knowledge forms in evaluating the quality of text sources is explored. Students viewed non-scientific sources as "poor" and scientific sources as "good," yet used both for information gathering. There were discrepancies between their assessment of source quality and actual use, indicating that perception of source quality doesn't always affect their practical decisions.
The third publication, Wellhöfer, Machleid, and Lühken (2023), investigates students' information practices in the lab, focusing on discourse between novice learners and experienced assistants. It shows that theoretical knowledge isn't sufficient for independent practical action, and students need actionable social information from experienced community members. The results highlight that information literacy in the lab for newcomers to a community of practice has distinctive features, and physical experience and tacit knowledge are crucial for learning the methods and group-specific knowledge of the practice community. The article demonstrates how learning information literacy in a practice community requires a social and physical experience and provides insights on how educators can support this process.
Mechanistic characterization of photoisomerization reactions in organic molecules and photoreceptors

In dieser Arbeit wurden verschiedene Einflüsse auf die Dynamik von Photoisomerisierungen in Phytochromen und indigoiden Photoschaltern untersucht. Beide Forschungsgebiete teilen wesentliche Aspekte wie die Kontrolle durch sterische Wechselwirkungen und den starken Einfluss der Polarität oder der ionischen Umgebung.
Auf dem Gebiet der Phytochrome wurde die relative Positionierung der knotenlosen Phytochrome innerhalb der Superfamilie der Phytochrome in Bezug auf ihre Photodynamik und den Effekt von Grundzustandsheterogenität herausgearbeitet. Es wurde anhand von ultraschnellen, zeitaufgelösten Anrege-Abtast-Experimenten der einzelnen GAF-Domäne All2699g1 im Vergleich mit dem vollständigen knotenlosen Phytochrom All2699g1g2 und dem strukturell ähnlichen knotenlosen Phytochrom SynCph2 gezeigt, dass knotenlose Phytochrome in ihrer Vorwärtsdynamik eine komplexe mehrphasige Kinetik mit einem langlebigen angeregten Zustand (~100 ps) aufweisen. Die beobachtete mehrphasige Kinetik konnte einer initialen Chromophordynamik sowie einer nicht exponentiellen Reorganisation der chromophor-umgebenden Proteinmatrix zugeordnet werden. Dies steht im starken Kontrast zur im Gebiet der Phytochrome etablierten Beschreibung derartiger mehrphasiger Kinetiken mittels heterogener Grundzustände. Stattdessen wurde ein konserviertes kinetisches Muster identifiziert, welches die mehrphasige Dynamik beschreibt und in allen in dieser Arbeit untersuchten Phytochrome beobachtet wurde. Zudem konnte dieses Muster in einem Phytochrom der Gruppe I und einem Phytochrom der Gruppe III, die einen ähnlichen Pr Dunkelzustand aufweisen, gezeigt werden, was eine breite Anwendbarkeit des damit verbundenen Mechanismus vermuten lässt. Weiterhin konnte die zentrale Rolle eines konservierten Tyrosins in der Photoisomerisierung anhand von Mutationsstudien in All2699g1 herausgearbeitet werden. Diese konservierte Aminosäure muss im Rahmen der Reorganisation der Proteinmatrix vom Chromophor weggezogen werden, damit die sterische Blockade abgebaut werden kann, die die Isomerisierung des Chromophors zunächst verhindert. Da diese Bewegung von diversen Faktoren in der den Chromophor umgebenden Proteinmatrix abhängt, weist sie eine nicht exponentielle Kinetik auf, die je nach Phytochrom, der spezifischen Flexibilität und dem vorhandenen Raum in der Bindetasche unterschiedliche Lebenszeiten aufweist.
Die Rückreaktion knotenloser Phytochrome konnte ebenfalls im Rahmen dieser Arbeit charakterisiert werden, welche im Pikosekundenbereich abläuft, und damit signifikant schneller ist als die Vorwärtsreaktion. Im Gegensatz zur Vorwärtsreaktion nimmt Grundzustandsheterogenität in der Rückreaktion eine weitaus bedeutendere Rolle ein. Hier weisen die in All2699g1 vorhandenen heterogenen Grundzustandspopulationen jeweils eine eigene Kinetik ihres angeregten Zustands auf, während die homogenen Grundzustände von All2699g1g2 und SynCph2 jeweils nur einen Zerfall des angeregten Zustands zeigen. Der Ursprung dieser Heterogenität konnte im Wasserstoffbrückennetzwerk des Chromophors lokalisiert und mit dem konservierten Tyrosin und einem konservierten Serin in der PHY-Domäne verknüpft werden. Die Anwesenheit der PHY-Domäne sorgt demnach für eine Verringerung der Grundzustandsheterogenität und des vorhandenen Raums in der Bindetasche, wodurch die Effizienz der Photoreaktion optimiert wird.
Zuletzt konnte die Millisekundendynamik knotenloser Phytochrome und der Einfluss der PHY-Domäne auf diese aufgeklärt werden. Die PHY-Domäne sorgt hierbei durch den verringerten Raum in der Bindetasche dafür, dass die zunächst stattfindende thermische Relaxation des Chromophors signifikant verlangsamt wird, während spätere Änderungen im Photozyklus nur wenig beeinflusst werden.
Auf dem Gebiet der indigoiden Photoschalter konnte, anhand eines sterisch überladenen Hemithioindigo Photoschalters, der Photoisomerisierungsmechanismus des Hula-Twists beobachtet und eine starke Lösungsmittelabhängigkeit der entsprechenden Kinetik aufgezeigt werden. Aus den durchgeführten zeitaufgelösten Anrege-Abtast-Experimenten in verschiedenen Lösungsmitteln konnte ein Modell für die Photodynamik des verwendeten Hemithioindigo Photoschalters entwickelt werden. In unpolaren Lösungsmitteln muss eine hohe Barriere zur produktiven konischen Durchschneidung überwunden werden, was zu Lebenszeiten des angeregten Zustands im Nanosekundenbereich führt. Der Weg zur produktiven konischen Durchschneidung folgt dabei dem Hula-Twist Mechanismus. Dieser Pfad ist in polaren Lösungsmitteln unerreichbar, weshalb eine schnelle Relaxation über eine unproduktive konische Durchschneidung stattfindet.
Im zweiten Projekt auf dem Gebiet der indigoiden Photoschalter wurde anhand der neuartigen Klasse der Iminothioindoxyl Photoschalter ein Schwingungsenergiedonor für Schwingungsenergietransferstudien entwickelt. Das daraus entwickelte Modellsystem, bestehend aus einer künstlichen Aminosäure auf Basis des Iminothioindoxyl Photoschalters und einem daran gekoppelten Schwingungsenergiesensor, wurde charakterisiert und die primäre Photoreaktion untersucht. Es konnte gezeigt werden, dass der angeregte Zustand des Modellsystems kurzlebig ist und unter Abgabe von großen Mengen an Schwingungsenergie zerfällt, unabhängig von der Anregungswellenlänge und dem verwendeten Lösungsmittel. Somit zeigt das entwickelte System vorteilhafte Eigenschaften für Schwingungsenergietransferstudien.
Insgesamt konnten somit die Mechanismen der Photoisomerisierungsreaktionen in knotenlosen Phytochromen und indigoiden Photoschaltern aufgeklärt und daraus die Relevanz der Umgebung für derartige Reaktionen herausgearbeitet werden.
"Oppenheimer" (Regie: Christopher Nolan, USA 2023) hat diverse Rekorde gebrochen. Er zählt zu den erfolgreichsten Filmen mit R-Rating; schon jetzt konnte er sich unter den ganz oder in wesentlichen Teilen vor dem Hintergrund des Zweiten Weltkriegs spielenden Filmen den vordersten Platz sichern. Das 180 Minuten lange Biopic über den sogenannten 'Vater der Atombombe' stellt selbst langjährige Spitzenreiter wie "Dunkirk" (Regie: Christopher Nolan, USA 2017) oder "Saving Private Ryan" (Regie: Steven Spielberg, USA 1998) in den Schatten. Mit einem erlesenen Star-Ensemble und einem Budget von 100 Millionen US-Dollar hat Nolan ein dunkles Historienspektakel geschaffen, das angesichts revolutionärer KI-Entwicklungssprünge, menschengemachter Klimaveränderungen und wiederaufkeimender geopolitischer Bedrohungen erschreckend aktuell ist. Wieder einmal sieht sich die Menschheit mit ihren selbstzerstörerischen Kräften konfrontiert. [...] "Oppenheimer" ist ein sehenswerter Film. Man sollte sich allerdings im Klaren darüber sein, dass er sich darin genügt, die Banalität einer Wissenschaft auszustellen, die im Krieg einfach zu funktionieren hat: Was gemacht werden kann, wird gemacht. Das Nachdenken darüber kommt - wie die Sichtbarkeit der Fortschrittskonsequenzen - immer erst ex post. Gerade mit Blick auf die historische Person Oppenheimer wird die gleichzeitig betriebene Mythisierung damit fragwürdig. Anstatt Oppenheimer zum modernen Prometheus, zum Vordenker amerikanischen Könnensbewusstseins zu stilisieren, hätte man mit gleichem Recht dessen weniger berühmten Bruder als mythische Vorlage wählen können. Schließlich war Epimetheus, der Nachdenker, dafür verantwortlich, dass die Büchse der Pandora in die Welt der Menschen kam.
Our mind has the function of representing the physical and social world we are in, so that we can efficiently interact with it. This results in a constant and dynamic interaction between mind and world that produces a balance when representations are at the same time accurate with respect to what the world is communicating to our organism, but also compatible with how our mind works.
A paradigmatic case of this interaction is offered by perception, which is the mental function that represents contingent aspects of the world built from what is captured by our senses. Indeed, the dominant philosophical view in cognitive science is that our perceptual states are representations of the world and not direct access to that world. These representational perceptual states therefor include the aspects of the world they represent and that initiate the perception by stimulating our sensory organs.
Perceptual representations are built using information from the sensory system, i.e., bottom-up information, but are also integrated with information previously acquired, i.e., top-down information, so that perception interacts with memory through language and other mental functions. Such organization is believed to reflect a general mechanism of our mind/brain, which is to acquire and use information to make efficient predictions about the future, continuously updating older information with present information.
This predictive processing works because the world is not random, but shows a regular structure from which reliable expectations can be built. One way that our minds make these predictions is by adapting to the structure of the world in an implicit, automatic and unconscious way, a process that has been called Implicit Statistical Learning (ISL). ISL is a learning process that does not require awareness and happens in an incidental and spontaneous way, with mere exposure to statistical regularities of the world. It is what happens when we learn a language during early childhood, and that allows us to be implicitly sensitive to the phonological structure of speech, or to associate speech patterns with objects and events to learn word meaning.
A specific case of ISL is the learning of spatial configuration in the visual world, which we apply to abstract arrays of items, but most importantly, also to more ecological settings such as the visual scenes we are immersed in during our everyday life. The knowledge we acquire about the structure of visual scenes has been called “Scene Grammar”, because it informs about presence and position of objects in a similar way to what linguistic grammar tells us about the presence and position of words. So, we implicitly acquire the semantics of scenes, learning which objects are consistent with a certain scene, as well as the syntax of scenes, learning where objects are positioned in a consistent way within a certain scene.
More recent developments have proposed that scene grammar knowledge might be organized based on a hierarchical system: objects are arranged in the scene, which offers the more general context, but within a scene we can identify different spatial and functional clusters of objects, called “phrases”, that offer a second level of context; within every phrase, then, objects have different status, with usually one object (“anchor object”) offering strong prediction of where and which are the other objects within the phrase (“local objects”). However, these further aspects of the organization of objects In scenes remain poorly understood.
Another problem relates to the way we measure the structure of scenes to compare the organization of the visual world with the organization in the mind. Typically, to decide if an object appears or not in a certain scene, and whether or not it appears in a certain position within a scene, researchers based their decision on intuition and common-sense, maybe validating those decisions with independent raters. But it has been shown that often these decisions can be limited and more complex information about objects’ arrangement in scenes can be lost.
A potential solution to this problem might be using large set of real-world images, that have annotations and segmentations of objects, to measures statistics about how objects are arranged in the environment. This idea exploits the nowadays larger availability of this kind of datasets due to increasing developments of computer vision algorithms, and also parallels with the established usage of large text corpora in language research.
The goals of the current investigation were to extract object statistics from this image datasets and test if they reliably predict behavioural responses during object processing, as well as to use these statistics to investigate more complex aspects of scene grammar, such as its hierarchical organization, to see if this organization is reflected in the organization of objects in our mind.
"A.I. or Nuclear Weapons: Can You Tell These Quotes Apart?" - so fragte die New York Times ihre Leser:innen am 10. Juni dieses Jahres. In Form eines metadiskursiven Ratespiels rückte die Zeitung damit eine Analogie in den Blick, die in den jüngsten Debatten um Künstliche Intelligenz zu erheblicher Prominenz gelangt ist. Wenn derzeit die Risiken neuester (Sprach-)Technologien beschworen werden, lässt der Vergleich mit dem Vernichtungspotenzial von Atomwaffen nicht lange auf sich warten. Dies gilt für die in Print- und Onlinemedien ausgetragene Diskussion, ist aber auch innerhalb der wissenschaftlichen Community mit ihren Spezialöffentlichkeiten zu beobachten. Warnungen vor den unabsehbaren Folgen der KI-Entwicklung gleichen in ihrer Drastik und teils bis aufs Wort den Mahnrufen und Appellen, mit denen in der Nachkriegszeit auf die damals neue Gefahr eines drohenden globalen Nuklearkonflikts reagiert wurde. Ob sich die von der New York Times anonymisiert präsentierten Aussagen wie "If we go ahead on this, everyone will die" oder "We are drifting toward a catastrophe beyond comparison" auf unsere Gegenwart oder auf die historische Situation der 1950er und 1960er Jahre beziehen, ist daher tatsächlich nicht immer ohne Weiteres auszumachen. Und erst kürzlich berichtete die Zeitung von einem Mitarbeiter des von Google betriebenen DeepMind-Forschungslabors, der einen Vortrag zum maschinellen Lernen mit einem Zitat aus George Orwells Essay "You and the Atomic Bomb" (1945) eröffnet hatte - ein weiteres Beispiel dafür, wie sehr die zwischen Künstlicher Intelligenz und Nuklearwaffen gezogenen Parallelen zu einem anscheinend unverzichtbaren Topos in der Auseinandersetzung mit neuronalen Netzen und Large Language Models (LLMs) geworden sind.
IL-38 is the latest discovered cytokine of the IL-1 family and has been added to the IL-36 subfamily. Since its discovery in 2001, increasing evidence suggests predominantly anti-inflammatory properties of IL-38, which are most likely exerted through three potential receptors, the IL-1 Receptor 1 (IL-1R1), IL-36 Receptor (IL-36R) and the IL-1 Receptor Accessory Protein Like 1 (IL-1RAPL1). However, to this date detailed knowledge of IL-38 functioning remains to be examined. Importantly, how IL-38 is processed, secreted from cells and the exact mechanisms of target receptor binding and intracellular signaling are not fully understood. Further, IL-38 has been associated with regulatory functions in autoimmune diseases like systemic lupus erythematosus (SLE) and psoriasis. At the same time however, connections between B cells as indispensable part of immunity and IL-38 remain rare.
In this study we examined the influence of IL-38 in peripheral human blood B cells differentiating into antibody secreting cells using a three-step in vitro differentiation process. We first show that all potential IL-38 binding receptors are present on peripheral blood B cells on a gene expression level and remain detectable throughout B cell differentiation. Next, while B cells treated with exogenous IL-38 depict no differences in early B cell activation markers, the process of B cell differentiation revealed significant alterations in B cell phenotype created by IL-38 treatment. Predominantly on day 7 of the differentiation process, IL-38 treated B cells showed significantly reduced CD38 expression which depicts an important step in development towards plasma cells. We hypothesize that IL-38 acts antagonistically on the IL-1R1 pathway reducing Nuclear factor kappa B (NFκB) expression and consequently decreasing CD38 expression. Further IL-38 reduced early antibody production while increasing IgM secretion at the end stages of differentiation. Next, we repeated the differentiation assays under the influence of additional IL-21 stimulation to further enhance plasma cell development. In these experiments, the impact of IL-38 on B cell differentiation and immunoglobulin production were reduced, indicating a comparatively moderate relevance of IL-38 for B cell differentiation. We then examined how proliferation and cell death were impacted by exogenous IL-38 during B cell differentiation. IL-38 treatment alone significantly reduced B cell survival which was further augmented by IL-21 stimulation. We conclude that IL-38 and IL-21 act synergistically in promoting B cell apoptosis, also depicting an anti-inflammatory property of IL-38. Finally, using a siRNA we successfully performed an IL-38 knockdown experiment of human blood B cells reducing IL-38 expression to 44% measured on day 4 of B cell differentiation. In these experiments we observed reversed tendencies of CD38 expression compared to exogenous IL-38 treatment. Here, IL-38 knockdown cells showed increased CD38 expression indicating endogenous regulatory properties of IL-38 in B cell differentiation.
Our project, for the first time proves direct effects of IL-38 on human B cells. The results support previous research of IL-38 to act anti-inflammatory as it seems to modulate B cell differentiation, survival, and immunoglobulin production in a down-regulatory manner. These findings pave way for more detailed research on the connection between B cell homoeostasis and IL-38 function.
The Heidelberg Ion-Beam Therapy Centre (HIT) provides proton, helium, and carbon-ion beams with different energies and intensities for cancer treatment and oxygen-ion beams for experiments. For several experiments and possible future applications, such as helium ion beam radiography, a low-intensity ion beam monitor integrated into the dose delivery feedback system for the accelerator control is a necessary pre-requisite. The updated 2D prototype for this purpose consists of scintillating fibres with enhanced radiation hardness, silicon photomultipliers (SiPMs) to amplify the emitted light, and a dedicated front-end readout system (FERS) to process and record the generated signals. This setup was tested successfully on monitoring ion-beam position and profile horizontally and vertically, as well as the beam intensity, for all four ion types with energies from 50 to 430 MeV/u and intensities from 1E2 to 1E7 ions/s. Additionally, time-of-arrival (ToA) measurements on single ions have been successfully performed for a limited intensity range, allowing for ion tracking in a further update. This will reduce noise, and will also improve the accuracy and usability of ion radiography.
The negative effect of fossil-based industrial processes on the environment, especially the contribution to global warming by emitting greenhouse gases such as CO2 causes a global threat to mankind. Therefore, technologies are demanded by the society for a sustainable and environmentally friendly economy. The biotechnological use of sugar-based feedstocks to produce valuable products are in conflict with, for example, food production. In order to overcome this issue, waste products such as syngas (H2, CO and CO2) or CO2 taken from the atmosphere are of increasing interest for biotechnological applications. Acetogenic bacteria are already used at industrial scale to produce sustainable and environmentally friendly biofuels from syngas. A promising candidate due to its physiological flexibility is the thermophilic acetogen Moorella thermoacetica. In contrast to most acetogens M. thermoacetica is not restricted to one energy conserving system. In addition to the Ech complex, cytochromes and quinones may be involved in energy conservation by, for example, DMSO respiration. The extra energy conserved can be used to form highly valuable but energy demanding products. In this review we give insights into the physiology of this acetogen, the current state of the art of M. thermoacetica as a platform for biotechnological applications and discuss future perspectives.
Affine Bruhat--Tits buildings are geometric spaces extracting the combinatorics of algebraic groups. The building of PGL parametrizes flags of subspaces/lattices in or, equivalently, norms on a fixed finite-dimensional vector space, up to homothety. It has first been studied by Goldman and Iwahori as a piecewise-linear analogue of symmetric spaces. The space of seminorms compactifies the space of norms and admits a natural surjective restriction map from the Berkovich analytification of projective space that factors the natural tropicalization map. Inspired by Payne's result that the analytification is the limit of all tropicalizations, we show that the space of seminorms is the limit of all tropicalized linear embeddings ι:Pr↪Pn and prove a faithful tropicalization result for compactified linear spaces. The space of seminorms is in fact the tropical linear space associated to the universal realizable valuated matroid.
Seit den 1970er Jahren haben sich die Universitäten für Senior*innen geöffnet; anfänglich nur durch die Öffnung bereits vorhandener Studiengänge, mehr und mehr aber auch durch gezielte Bildungsangebote für die Zielgruppe Älterer. Die „Universität des 3. Lebensalters“ (U3L) an der Universität Frankfurt bietet seit 40 Jahren Angebote für Ältere und hat daher Einblicke in die Bildungsbedürfnisse dieser Zielgruppe im Wandel der Zeit gesammelt: Überwog früher das Nachholen akademischer Bildung als Motiv für die Teilnahme, so geht es den sogenannten „Babyboomern“ (Jahrgänge 1955-1964) vielfach um Selbstreflexion von Lebensgestaltung und Lebensvollzug. Am Beispiel der beiden mehrjährigen Lehr-/Lern-Projekte „Biografie und Gesellschaft“ und „Männlichkeiten im Altern“ zeigt der Autor im vorliegenden Beitrag, wie eine teilnehmer*innenorientierte Seminargestaltung für diese Zielgruppe didaktisch erfolgen kann: die Inhalte in allen Phasen der didaktischen Planung entlang der biografisch begründeten Interessen der Teilnehmer*innen ausrichten, die Seminare partizipativ anlegen, eigene und fremde Erfahrungen einbeziehen, Emotionen achtsam aufgreifen und reflektieren. Als Lehrender beschreibt er sich als neugieriger und interessierter Ermöglicher und Begleiter von Lernprozessen, der sich auch selbst als alternder Mit Mensch einbringt.
Homeownership rates differ widely across European countries. We document that part of this variation is driven by differences in the fraction of adults co-residing with their parents. Comparing Germany and Italy, we show that in contrast to homeownership rates per household, homeownership rates per individual are very similar during the first part of the life cycle. To understand these patterns, we build an overlapping-generations model where individuals face uninsurable income risk and make consumption-saving and housing tenure decisions. We embed an explicit intergenerational link between children and parents to capture the three-way trade-off between owning, renting, and co-residing. Calibrating the model to Germany we explore the role of income profiles, housing policies, and the taste for independence and show that a combination of these factors goes a long way in explaining the differential life-cycle patterns of living arrangements between the two countries.
Biotechnological processes offer better production conditions for a wide variety of goods of industrial interest. The production of aromatic compounds, for example, involves molecules of great value for cosmetic, plastic, agrochemical and pharmaceutic industries. However, the yield of such processes frequently prevents a proper implementtation that would allow the replacement of traditional production processes.
Numerous rational engineering approaches have been attempted to enhance metabolic pathways associated with desired products. Unfortunately, genetic modifications and heterologous pathway expression often lead to a higher metabolic burden on the producing organisms, ultimately leading to reduced production levels and fitness.
This project utilised adaptive laboratory evolution to better understand the development of synthetic cooperative consortia, using S. cerevisiae as a model organism. Specifically, a synthetic cooperative consortium was developed around the exchange of lysine and tyrosine, which was subjected to adaptive laboratory evolution aiming to induce mutations that would improve the system’s fitness either by enhanced production or upgraded stress resistance. Consequently, the mutant strains isolated after the evolution rounds were sequenced to identify relevant variations that could be related to the growth and production phenotypes observed.
The insights derived from this project are expected to contribute to further developing synthetic cooperative consortia with utilitarian purposes.
Methods using environmental DNA to explore and analyze biodiversity from previously unexplored habitats and ecosystems have become increasingly popular in recent years. This is particularly due to the potential reduction in necessary taxonomic expertise, the opportunity to assess microorganismal communities, and decreased time investments required to cover large spatial extents. In forests, the surface of tree bark is an important habitat for epiphytic diversity. Because of the large surface area rich in micro-niches, the seasonal stability of the substrate, and the longevity of trees, tree bark surfaces provide an ideal habitat for many species. Yet, we lack a comprehensive understanding of their communities and the environmental drivers behind the community assembly. These missing links hinder the exploration of the forest microbiome as a whole and limits our understanding of functions of a large forest habitat and its connections to other forest microbiomes. With a holistic eDNA metabarcoding approach, encompassing samples of three major taxonomic groups (e.g. bacteria, fungi, and green algae), as well as simultaneous collections from multiple forest habitats we can contribute to closing these gaps and increase our knowledge of the forest microbiome.
My dissertation is set within the framework of the Biodiversity Exploratories and was conducted in four parts: I. the establishment of an eDNA metabarcoding workflow to reveal the local diversity of the bark surface microbiome; II. the upscaling of the method to large geographic and environmental gradients to uncover the drivers of the microbiome; III. the integration of soil and bark samples to investigate compositional differences in two important forest habitats; IV. the evaluation of eDNA metabarcoding as a tool for biodiversity assessments of lichen diversity in forests.
In the first part, I developed a simple, cost-effective and fast sampling strategy to acquire eDNA samples from the bark of trees in forest ecosystems. Using readily available medical-specimen-collection swabs I sampled bark surfaces of individual trees in Central German forests and used metabarcoding to amplify marker genes of green algae, fungi and bacteria. From the sequencing reads I calculated the first diversity estimates of the major organismal groups of bark surface microbiomes from Central European forests. Overall the methodology produced reliable results, allowing for an expanded sampling in the second part.
In the second part of the dissertation, I expanded the sampling based on the results of part one. I collected bark surface samples from the three regions of the Biodiversity Exploratories covering large spatial and environmental gradients representative for Central European forests. The collection included composite samples from 150 plots and over 750 trees. Utilizing measurements of climatic and forest structure variables provided by the Biodiversity Exploratories, as well as my own community data, I identified the biotic and abiotic drivers behind alpha and beta diversity of the bark surface microbiome.
In the third part, I studied the differences between the bark surface as an unexplored and the soil as an example of a well characterized forest microbiome. Using only the fungal part of the large sampling campaign and soil samples obtained from the same plots at the same time, I assessed the commonalities and differences of the micro-communities of these distinct forest niches. Furthermore, I included two coniferous and one deciduous tree species to examine, if the effect of tree species, previously shown for soil microbiomes, also holds true for the bark surface.
In the last part of my dissertation, I used eDNA in a more applied way as a tool in biodiversity assessments of lichenized fungi. I compared the results from eDNA metabarcoding to an expert floristic mapping conducted in the same plots in 2007/2008. I assigned functional guilds to the fungal taxa obtained in the large sampling campaign and used a subset that was assigned as lichenized fungi.
In conclusion, I showed that eDNA metabarcoding is a valuable tool to reveal the unknown diversity of microorganisms in forest ecosystems. In particular, my results advance our understanding of the bark surface microbiome, an underexplored habitat within forests. The tightly linked interactions of the three major microbial groups underline that studies need to take holistic approaches across multiple taxonomic groups to deepen our understanding of processes governing the assembly of microbiomes. Results from my dissertation may serve as a foundation to inform hypotheses addressing the functions of forest microbiomes. The massive diversity data collected may also contribute to closing the gap in our understanding of macro-organisms and micro-organisms with respect to diversity distributions and patterns of richness, and serve as a baseline for predictions of biodiversity responses under future anthropogenic change.
Untersuchung der Expression von Wachstumsfaktoren in reseziertem Hirngewebe von Epilepsiepatienten

Hintergrund: Die Epilepsie gehört zu den häufigsten chronischen neurologischen Erkrankungen beim Menschen. Bei Patienten mit mesialer TLE und Hippocampussklerose besteht die höchste Wahrscheinlichkeit, eine medikamentöse Therapierefraktärität zu entwickeln. Die Ursache der Hippocampussklerose sowie die ursächlichen Mechanismen sind nicht bekannt. Allerdings kann eine initiale Schädigung, wie etwa komplizierte Fieberkrämpfe im Kindesalter, Schädel-Hirn-Traumata, Schlaganfälle, entzündliche Prozesse oder Ähnliches, für die Entwicklung einer Hippocampussklerose prädisponieren. Diese kann anschließend nach einer klinisch stummen Latenzperiode zur Entwicklung spontaner epileptischer Anfälle und der Diagnose einer Epilepsie führen. Im Rahmen der Epileptogenese, also der Entstehung und Progression der Epilepsie kommt es zu Wachstumsprozessen, weshalb eine Beteiligung von neurotrophen Wachstumsfaktoren naheliegend war. Das Ziel dieser Arbeit war die vergleichende Untersuchung resezierter Hippocampi auf Wachstumsfaktoren, um semiquantitative Daten zu deren Verteilung bei Epilepsiepatienten zu erhalten. Des Weiteren war die Korrelation mit den klinischen Daten der Patienten von besonderem Interesse, da so Hinweise auf mögliche Zusammenhänge zwischen dem klinischen Erscheinungsbild und der Expression der Wachstumsfaktoren gewonnen werden konnten.
Methoden: Bei dem in der vorliegenden Arbeit untersuchten Gewebe handelt es sich um Hippocampi von 21 Patienten mit TLE, die epilepsiechirurgisch therapiert wurden. Die Schnitte der paraffinierten Hippocampi wurden mittels Immunhistochemie auf die Wachstumsfaktoren BDNF, FGF2, GDNF, GMFB und PDGF-B untersucht. Im Anschluss wurden die Schnitte gescannt und die Zellen mittels eines Algorithmus identifiziert und ausgewertet. Diese experimentellen Daten wurden anschließend mit den klinischen Daten der Patienten korreliert.
Ergebnisse: Es fand sich eine signifikante Korrelation zwischen der Expression von GMFB und dem postoperativen Outcome der Patienten. Des Weiteren fanden sich auch Korrelationen zwischen der präoperativen Anfallsfrequenz und der Expression von BDNF sowie GDNF. Auch die Epilepsiedauer korrelierte mit der Expression von BDNF. Zudem fanden sich Korrelationen zwischen den Ergebnissen der neuropsychologischen Testungen und der Expression von BDNF, sowie PDGF-B.
Diskussion: Die vorliegende Arbeit liefert einige Daten, die Hinweise für nachfolgende Untersuchungen geben können. Sowohl für die Anfallsfrequenz, als auch für die Epilepsiedauer fanden sich signifikante Korrelationen mit BDNF. Beides ist passend zu den vermuteten und zum Teil in der Literatur beschriebenen Mechanismen im Rahmen der Epilepsie, also einer postiktalen Hochregulation von Wachstumsfaktoren beziehungswiese des Zugrundegehens von Zellen im Verlauf der Erkrankung und damit zu einer reduzierten Expression von Wachstumsfaktoren. Geschlechterabhängige Unterschiede in der Expression der Wachstumsfaktoren fanden sich, passend zu der vorhandenen Literatur, nicht. Interessant ist, dass sowohl Geschlechtshormone als auch anfallssuppressive Medikamente einen Einfluss auf die Expression der Wachstumsfaktoren haben können.
Bis heute ist kein Biomarker bekannt, der eine Vorhersage über den Erfolg einer operativen Therapie bei therapierefraktären TLE treffen kann. Da meine Daten eine Korrelation von GMFB und dem postoperativen Outcome zeigen, bietet es sich für weitere Untersuchungen an, GMFB als präoperativen Biomarker zu nutzen. zu können, wäre eine einfachere Probengewinnung beispielsweise aus Blut, Liquor, Urin oder Speichel notwendig. Im Sinne einer „Liquid Biopsy“ könnte so der Erfolg einer chirurgischen Therapie weiter objektiviert werden, was die Entscheidungsfindung einfacher und risikoärmer gestalten würde.
Background and purpose: In patients with epilepsies of structural origin, brain atrophy and pathological alterations of the tissue microstructure extending beyond the putative epileptogenic lesion have been reported. However, in patients without any evidence of epileptogenic lesions on diagnostic magnetic resonance imaging (MRI), impairment of the brain microstructure has been scarcely elucidated. Using multiparametric quantitative (q) magnetic resonance imaging MRI, we aimed to investigate diffuse impairment of the microstructural tissue integrity in MRI-negative focal epilepsy patients.
Methods: 27 MRI-negative patients with focal epilepsy (mean age 33.1 ± 14.2 years) and 27 matched healthy control subjects underwent multiparametric qMRI including T1, T2, and PD mapping at 3 T. After tissue segmentation based on synthetic anatomies, mean qMRI parameter values were extracted from the cerebral cortex, the white matter (WM) and the deep gray matter (GM) and compared between patients and control subjects. Apart from calculating mean values for the qMRI parameters across the respective compartments, voxel-wise analyses were performed for each tissue class.
Results: There were no significant differences for mean values of quantitative T1, T2, and PD obtained from the cortex, the WM and the deep GM between the groups. Furthermore, the voxel-wise analyses did not reveal any clusters indicating significant differences between patients and control subjects for the qMRI parameters in the respective compartments.
Conclusions: Based on the employed methodology, no indication for an impairment of the cerebral microstructural tissue integrity in MRI-negative patients with focal epilepsy was found in this study. Further research will be necessary to identify relevant factors and mechanisms contributing to microstructural brain tissue damage in various subgroups of patients with epilepsy.
Die chronische myeloische Leukämie (CML) ist eine klonale myeloproliferative Neoplasie und hat ihren Ursprung in transformierten pluripotenten Stammzellen im Knochenmark. Der Krankheitsentstehung liegt eine reziproke chromosomale Translokation zugrunde, in deren Folge ein neues Fusionsgen, das sogenannte Philadelphia-Chromosom, entsteht. Das hiervon codierte Genprodukt ist eine Tyrosinkinase mit konstitutiver Aktivität mit resultierender unkontrollierter Signaltransduktion. Gegen diese Tyrosinkinase existiert eine molekular zielgerichtete Therapie, die Tyrosinkinase-Inhibitoren (TKI).
Den Therapiestandard stellt die Behandlung mit einem TKI dar. Seit einigen Jahren existiert jedoch das Therapieziel der therapiefreien Remission (TFR), bei dem CML-Patienten mit einem guten molekularen Ansprechen den TKI nach einiger Zeit absetzen und unter engmaschigen Kontrollen therapiefrei bleiben. Ungefähr die Hälfte dieser Patienten erleidet kein molekulares Rezidiv und bleibt langfristig in TFR. Zu der Thematik der TFR existieren zahlreiche klinische Studien, die die Umsetzbarkeit und die Sicherheit eines Absetzversuchs belegen und Kriterien definiert haben, die für einen Absetzversuch erfüllt sein sollten. In dieser Dissertation wird die Umsetzung der TFR im klinischen Alltag onkologischer Praxen untersucht. Es wird untersucht, ob die Studienergebnisse mit den TFR-Raten und den identifizierten Einflussfaktoren auf den Praxisalltag übertragbar sind und ob das Therapieziel TFR in den klinischen Alltag integrierbar ist. Hierfür werden die Daten von 61 CML-Patienten mit einem Absetzversuch aus fünf onkologischen Praxen retrospektiv ausgewertet. Erhoben werden Parameter der Routineversorgung und die Ergebnisse der Kontrolluntersuchungen während der TFR werden dokumentiert und ausgewertet. Die TFR-Raten von ca. 50%, die in den Absetz-Studien beobachtet wurden, finden sich im hier untersuchten Patientenkollektiv wieder. Mithilfe der binären logistischen Regression wird getestet, ob bestimmte Faktoren, wie die Therapiedauer und das verwendete Medikament vor dem Absetzen, einen signifikanten Einfluss auf den Verlauf der TFR haben. Hier kann kein signifikanter Einflussfaktor identifiziert werden. Es lässt sich eine signifikant längere therapiefreie Überlebensdauer bei den Patienten zeigen, die mit einem TKI der zweiten Generation vor dem Absetzen behandelt wurden.
Zusammenfassend lässt sich sagen, dass die TFR bei geeigneten CML-Patienten in der Praxis umsetzbar und sicher ist und fest im klinischen Alltag onkologischer Praxen verankert sein sollte.
Meliolales (black mildews) is an order of plant parasitic ascomycetous fungi in the tropics and subtropics. They are frequently overgrown and parasitized by other fungi, known as hyperparasites. During the last few years, species of hyperparasitic fungi on Meliolales have been collected in Benin and Panama. A new species of Paranectria and seven new reports of hyperparasites of different systematic groups are presented here with detailed descriptions and illustrations, together with new data concerning fungal hosts and host plants. The new species is called Paranectria longiappendiculata, characterized by exceptionally long appendages carried by the ascospores. New records for Benin and Panama are Calloriopsis herpotricha, Dimerosporiella cephalosporii, Isthmospora glabra, Isthmospora trichophila, Malacaria meliolicola, Paranectriella hemileiae, and Paranectriella minuta. Calloriopsis herpotricha is recorded for Africa and D. cephalosporii and P. hemileiae for America for the first time, suggesting an apparently pantropical distribution. Findings show a blatant lack of investigation on hyperparasitic fungi in the tropics. The phylogenetic positions of three of these newly reported species, C. herpotricha, D. cephalosporii, and P. minuta, are shown based on the analysis of internal transcribed spacer (ITS), large subunit (LSU), and small subunit (SSU) rDNA sequences. These sequences were generated in the context of the present study for the first time.
According to the inflationary theory of cosmology, most elementary particles in the current Universe were created during a period of reheating after inflation. In this Letter, we self-consistently couple the Einstein-inflaton equations to a strongly coupled quantum field theory as described by holography. We show that this leads to an inflating universe, a reheating phase, and finally a universe dominated by the quantum field theory in thermal equilibrium.
In narratology, a widely recognized method involves exploring the connection between implied authors and implied readers. It entails correlating abstract narrative components within a text to understand the conveyed message and the multitude of interpretations it can offer. The present study adopts an implied reader-oriented approach to analyze three selected novels from the twentieth and twenty-first centuries—one Nigerian, one Caribbean, and one Kurdish. The aim is to explore the potential readings within these texts, considering the hermeneutic process of critical reading. The selected texts include Chinua Achebe’s Things Fall Apart, (1958), Same Selvon’s The Lonely Londoners, (1956), and Karwan Kakesur’s The Channels of the Armed Monkeys, (2011). This approach closely examines the communication between the author and reader of the text, with a special focus on the varying levels of communication between the components of the narration, including fictional and implied fictional communication.
The implied fictional communication occurs between a narrative agent known as ‘the implied author’ and its fictional counterpart ‘the implied reader’ rather than between the real, flesh and blood authors and readers. I argue that this level of communication is coded, and the act of decoding it is part of the reading process performed by the reader. Certain texts can propose different and sometimes opposing readings which are initially and purposefully designed by the implied author and addressed to different implied readers. These readings are not necessarily the results of different real readers but rather incorporated ones predetermined by the implied author only to be acknowledged and uncovered by the readers. In other words, the latent meaning is and always was an integral part of the text and is not something created by the imaginative reader or critic. The core interest of my thesis lies in identifying prompts and suggestions within the narrative of the selected texts and ultimately understanding the readerships prestructured in them. Identifying the different readers within those texts will provide new reinterpretations that can add undetected values to the reading process and sometimes suggests opposing readings to how those texts have so far been read. Additionally, it is the objective of this thesis to propose new ways that readers can interact with reading literature that would result in a more aesthetic and entertaining reading experience besides providing ways to be more informed and aware of the cues certain narrative texts contain.
There have been numerous critical studies on both narratology and postcolonial or minority literatures; however, there has been little scholarly work that attempts to utilize narratology as a theoretical foundation for understanding postcolonial and minority fiction.
This study examines fictional texts from Nigerian, Caribbean, and Kurdish literature, employing the narratological concept known as ‘Multiple Implied Readers’. By incorporating concepts from Brian Richardson’s ‘Singular Text, Multiple Implied Readers’, and Peter J. Rabinowitz’s ‘authorial audiences’, I explore the various readerships that the texts could encompass. This exploitation may lead to the discovery of new readings, interpretations, and meanings that would otherwise remain undetected. These structures introduce provocative indeterminacies that challenge the reader’s synthesis of information into coherent configurations of meaning. Consequently, this approach not only enhances the reading experience but also opens doors to new interpretations of the text. In some cases, these interpretations could even dismantle prior understandings and propose entirely new readings.
The concepts of the implied author and implied reader have been studied before in relation to various disciplines of narratology. However, by applying them in conjunction with the relatively less researched subject of multiple implied readers, I aim to shed light on important aspects of these readings. This exploration could prove beneficial for literature students as well as critical readers of literary texts, revealing the potential of these texts to accommodate more than one implied reader within their narratives.

In der vorliegenden Arbeit wurden die Proteine Vlp15/16 und GlpQ aus B. miyamotoi hinsichtlich ihrer Eigenschaft, mit Plasminogen zu interagieren, charakterisiert.
Da einige Fälle von ZNS-Beteiligungen bei B. miyamotoi-Infektionen berichtet wurden, ist anzunehmen, dass diese Borrelienspezies über molekulare Mechanismen zur Überwindung der Blut-Hirn-Schranke verfügt. Eine solche Strategie könnte die Bindung wirtseigener Proteasen wie z.B. Plasminogen sein, um Komponenten der extrazellulären Matrix zu degradieren und dadurch die Dissemination des Erregers zu erleichtern.
Während Vmps, zu welchen auch Vlp15/16 gehört, als membranständige Proteine durch Variation der antigenen Oberflächenmatrix zur Immunevasion des Erregers beitragen, ist GlpQ bei der Hydrolyse von Phospholipiden in den Zellstoffwechsel eingebunden. Trotz dieser unterschiedlichen Funktionen, die den beiden Proteinen zukommen, binden beide Moleküle Plasminogen. Die Eigenschaften dieser Interaktion wurden in dieser Arbeit im Detail untersucht. Die Ergebnisse zeigen, dass Vlp15/16 und GlpQ Plasminogen konzentrationsabhängig binden und die Dissoziationskonstanten (Vlp15/16:Kd = 354 nM ± 62 nM; GlpQ: Kd = 413 nM ± 72 nM) für beide Proteine im Bereich der Serumkonzentration von 2 µM liegen. Darüber hinaus konnte gezeigt werden, dass den beiden Proteinen unterschiedliche Mechanismen zugrunde liegen, Plasminogen zu binden. Während die erhobenen Daten für Vlp15/16 darauf hindeuten, dass Lysin-Reste essenziell für die Interaktion sind, scheinen bei GlpQ ionische Wechselwirkungen von Bedeutung zu sein.
Um die Beteiligung von C-terminal lokalisierten Lysin-Resten für die PlasminogenBindung von GlpQ nachzuweisen, wurden Varianten mit einzelnen Lysin-Substitutionen an zwei unterschiedlichen Positionen (333 und 334) sowie eine Variante mit einer Zweifach-Substitution (GlpQ-K333A-K334A) generiert. Die Bindungsanalysen ergaben, dass insbesondere der Lysin-Rest an Position 334 bei der Interaktion mit Plasminogen beteiligt ist.
Die funktionellen Analysen zeigten, dass das an Vlp15/16 beziehungsweise GlpQ gebundene Plasminogen zu Plasmin aktiviert werden konnte und darüber hinaus dazu in der Lage war, das physiologische Substrat Fibrinogen zu degradieren.
Abschließend wurde die Plasminogen-Bindung an nativen B. miyamotoi-Zellen mittels Immunfluoreszenz-Mikroskopie nachgewiesen.
Die Ergebnisse dieser Arbeit weisen Vlp15/16 und GlpQ als Plasminogen-bindende Proteine aus, mit deren Hilfe B. miyamotoi befähigt ist, Komponenten der extrazellulären Matrix zu degradieren und somit prinzipiell zur Dissemination des Erregers beizutragen.
The introduction informs about Black literary imaginations of Europe that reverse or complicate the (neo-)colonialist European gaze at the "African Other". It reviews the state of research and provides an overview of the aims and sources of the special issue, whose individual contributions take into account both national specificities and transnational contexts. Sandra Folie and Gianna Zocco emphasise the important role of comparative literature for the field of African European studies (and vice-versa).
Abdominal aortic aneurysm (AAA) is the most common type of aortic aneurysm, which is defined as a dilation of the abdominal aorta over 3.0 cm or more. Surgical repair is the golden standard for the treatment of AAA, in which open surgical repair (OSR) and endovascular aneurysm repair (EVAR) are the main approaches. Technically speaking, the lesion segment of aueurysm is completely replaced by a graft during OSR, while in EVAR, the lesion is insulated by a stentgraft. EVAR is a less invasive treatment than OSR and shows a lower early mortality rate, although the long-term advantages of EVAR over OSR remain inconclusive.
Endoleak, especially the type II endoleak (T2EL), is a common complication after EVAR. According to research, 16-28% of the patients develop a T2EL after EVAR, and it accounts for nearly three in four of all types of endoleaks. Around 30-50% of the T2EL resolved spontaneously during the follow-up, however, it still causes a secondary intervention in many patients. Therefore, it is critical to monitor endoleaks after repair.
Patent aortic branches in the stent-overlapped area and vasa vasorum have been identified as potential sources of blood flow in T2EL. However, the mechanisms of biological changes or remodeling of the aneurysm sac after the repair are still not clear, but they have been considered to play an important role in the development of endoleaks. Unfortunately, it is impossible to obtain a tissue sample of the aortic wall in patients who underwent EVAR.
MicroRNAs (miRNAs) are a class of small single-stranded non-coding RNAs that inhibit the expression of target message RNA (mRNA). miR-29b/29c, miR-155, and miR-15a are miRNAs associated with regulating extracellular matrix (ECM) components, inflammation, and proliferation, respectively. All four miRNAs have been identified as biomarkers of AAA, not only in aneurysm tissue but also extracellular as circulating miRNAs. However, it is still unknown whether they can reflect the biological changes after AAA repair. Thus, we conducted a prospective study to investigate the changes in expression of circulating miR-29b, miR-29c, miR-155, and miR-15a before (T0), 3 days (T1), and 3 months (T2) after AAA repair.
A total of 39 patients were recruited for this study, 17 of whom were repaired by OSR and 22 of whom were repaired by EVAR. Four patients failed the T2 follow-up due to the Covid-19 pandemic. No significant changes were found in the expression of miR-29b, miR-29c, miR-155, and miR-15a. There were also no obvious differences between OSR and EVAR. However, the T1 expression of miR-15a was significantly lower in patients without endoleak after EVAR than in those who developed endoleak after EVAR and those who were repaired by OSR. Unfortunately, these differences did not persist to the T2 follow-up, and no other differences were found among these patients.
In summary, miR-15a is a miRNA that significantly changes in AAA patients. This study demonstrates that the expression of circulating miR-15a is lower in patients without endoleak three days after EVAR, compared to those who had endoleak after EVAR and those who underwent OSR. The results suggest that miR-15a might be involved in the early aortic remodeling after EVAR as an indicator of endoleak.
Human functional brain connectivity can be temporally decomposed into states of high and low cofluctuation, defined as coactivation of brain regions over time. Rare states of particularly high cofluctuation have been shown to reflect fundamentals of intrinsic functional network architecture and to be highly subject-specific. However, it is unclear whether such network-defining states also contribute to individual variations in cognitive abilities – which strongly rely on the interactions among distributed brain regions. By introducing CMEP, a new eigenvector-based prediction framework, we show that as few as 16 temporally separated time frames (< 1.5% of 10min resting-state fMRI) can significantly predict individual differences in intelligence (N = 263, p < .001). Against previous expectations, individual’s network-defining time frames of particularly high cofluctuation do not predict intelligence. Multiple functional brain networks contribute to the prediction, and all results replicate in an independent sample (N = 831). Our results suggest that although fundamentals of person-specific functional connectomes can be derived from few time frames of highest connectivity, temporally distributed information is necessary to extract information about cognitive abilities. This information is not restricted to specific connectivity states, like network-defining high-cofluctuation states, but rather reflected across the entire length of the brain connectivity time series.
An important question concerning inter-areal communication in the cortex is whether these interactions are synergistic, i.e. brain signals can either share common information (redundancy) or they can encode complementary information that is only available when both signals are considered together (synergy). Here, we dissociated cortical interactions sharing common information from those encoding complementary information during prediction error processing. To this end, we computed co-information, an information-theoretical measure that distinguishes redundant from synergistic information among brain signals. We analyzed auditory and frontal electrocorticography (ECoG) signals in five common awake marmosets performing two distinct auditory oddball tasks and investigated to what extent event-related potentials (ERP) and broadband (BB) dynamics encoded redundant and synergistic information during auditory prediction error processing. In both tasks, we observed multiple patterns of synergy across the entire cortical hierarchy with distinct dynamics. The information conveyed by ERPs and BB signals was highly synergistic even at lower stages of the hierarchy in the auditory cortex, as well as between auditory and frontal regions. Using a brain-constrained neural network, we simulated the spatio-temporal patterns of synergy and redundancy observed in the experimental results and further demonstrated that the emergence of synergy between auditory and frontal regions requires the presence of strong, long-distance, feedback and feedforward connections. These results indicate that the distributed representations of prediction error signals across the cortical hierarchy can be highly synergistic.

Mutoni im Un/Happyland : die Bürde weißer Retter*innen in Tete Loepers Roman "Barfuß in Deutschland"

In Tete Loeper's novel "Barefoot in Germany" (2020), Black first-person narrator Mutoni from Rwanda recounts her experiences as a marriage migrant, sex worker, maid, and caregiver in Germany, a supposed "Happyland" where racism is considered the offense of "others": bad individuals and Nazis. However, Loeper's white savior characters are both nice people and (unwitting) racists, while some of Mutoni's Black sisters behave in discriminatory ways as well. Drawing on critical race theory and imagology, this article shows how the novel deconstructs and appropriates stereotypical images from "'colorblind' Europe" on both a thematic and formal-aesthetic level. By engaging with a comparative and transnational frame of reference that goes beyond a monolingual white canon of theory and literature, the article reveals the novel's connections to other Black texts and genres, as well as its literary strategies in dealing with identity (politics).
ADHS im Erwachsenenalter ist eine komplexe Erkrankung, die durch verschiedene diagnostische Instrumente erfasst wird. Aktuell spielen dabei vor allem die klinische Exploration, Interview- und Fragebogendaten eine Rolle.
Obwohl bei Patient*innen vielfältige kognitive Einschränkungen bestehen, wird die Verwendung neuropsychologischer Daueraufmerksamkeitstests (CPTs) in den klinischen Leitlinien nur ergänzend erwähnt. Validität und Nützlichkeit von CPTs bei ADHS Patient*innen wird aufgrund der mangelnden Sensitivität und
Spezifität immer wieder kritisch diskutiert. Multidimensionale Ansätze, in denen verschiedene neuropsychologische Beurteilungen mit klinischen Parametern kombiniert werden, sollen das Aufstellen einer ADHS Diagnose im Erwachsenenalter verbessern. Die vorliegende Studie untersucht zwei verschiedene CPTs bezüglich deren Vergleichbarkeit und deren Verknüpfung mit klinischen Parametern. Ein direkter Vergleich innerhalb derselben Stichprobe ist bisher nicht durchgeführt worden, könnte jedoch wichtige Implikationen für die Testauswahl und darauffolgende klinische Interventionen haben. Die klinische Stichprobe besteht aus zufällig ausgewählten erwachsenen Patient*innen, die sich in der ADHS Ambulanz der Klinik für Psychiatrie, Psychosomatik und Psychotherapie des Universitätsklinikums Frankfurt zur diagnostischen ADHS Beurteilung zwischen Mai und November 2019 vorstellten. Die Patient*innen wurden von erfahrenen Ärzt*innen mithilfe des strukturierten diagnostischen Interviews für ADHS bei Erwachsenen (DIVA 2.0) befragt. Au§erdem wurden Schulzeugnisse bewertet und die medizinische Vorgeschichte erhoben. Anschließend wurden die Patient*innen mit dem Wender-Reimherr Interview (WRI) von einer unabhängigen Interviewerin befragt und füllten die folgenden Selbstbeurteilungsfragebögen aus: Kurzform der Wender-Utah Rating Scale (WURS-k), Impulsive Behavior Scale (UPPS) und General Health Questionnaire (GHQ-28). Alle Patient*innen nahmen anschließend an der Testung mit zwei CPTs teil: dem Quantified Behavior Test (Qb-Test) und Nesplora Aquarium [9]. Der Qb-Test misst neben Unaufmerksamkeit und Impulsivität auch Hyperaktivität mithilfe einer Infrarot Kamera und eines Reflektors, welche das Bewegungsmuster während der Testung aufzeichnen. In Nesplora Aquarium wird virtuelle Realität genutzt, um das Bewegungsmuster mithilfe eines optischen Gerätes, das mit Sensoren und Lautsprechern ausgestattet ist, aufzuzeichnen. Es zeigte sich, dass die übergreifende Vergleichbarkeit beider Tests begrenzt ist. Trotz der kongruenten Messung von Unaufmerksamkeit in beiden Tests konnten keine Korrelationen mit den klinischen Parametern festgestellt werden. Hyperaktivität ist in beiden Tests positiv mit aktuellen ADHS Symptomen assoziiert, wobei der Qb-Test darüber hinaus noch Korrelationen mit ADHS Symptomen aus der Kindheit aufweist. Impulsivität stellte sich als ein unabhängiges Konstrukt heraus, sowohl die Korrelation zwischen beiden Tests als auch die Assoziation mit klinischen Parametern betreffend. Dies stellt in Frage, ob ein homogenes mpulsivitätskonstrukt existiert, welches sich in neuropsychologischen Tests und klinischen Symptomen abbildet. Insgesamt ermöglicht die Studie einen guten Vergleich beider CPTs und zeigt detaillierte Informationen über die
Kompatibilität mit klinischen Symptomen auf. Unaufmerksamkeit stellte sich als ein basaler Faktor heraus, der zwischen beiden Tests über alle Patient*innen hinweg überlappt. Da dieser Faktor jedoch nicht mit klinischen Parametern korreliert, unterstreicht dies die Problematik, subjektive mit objektiven Methoden in der ADHS Diagnostik zu verbinden. Die aktuellen Ergebnisse bezweifeln die einfache Anwendung sogenannter objektiver Messverfahren in der klinischen ADHS Diagnostik aufgrund ihrer fragwürdigen Validität. Die Ergebnisse der Studie verdeutlichen die Notwendigkeit eines neuen diagnostischen Ansatzes im Sinne der Präzisionsmedizin, welcher über die konventionellen Klassifikationssysteme hinaus passende individuelle Lösungen für Diagnostik und Behandlung ermöglicht.
In der vorliegenden Dissertation wird auf diesen Ergebnissen basierend der Goldstandard der aktuellen ADHS-Diagnostik unter Beachtung der Leitlinien diskutiert. Die untersuchten CPTs werden unter Berücksichtigung der Gütekriterien guter diagnostischer Verfahren überprüft. Das Potenzial neuropsychologischer Testverfahren im Hinblick auf die Steigerung von Objektivität, Validität und Differenziertheit der Diagnosen wird untersucht und Empfehlungen für die klinische Anwendung formuliert.
The mitochondrial matrix peptidase CLPP is crucial during cell stress. Its loss causes Perrault syndrome type 3 (PRLTS3) with infertility, neurodegeneration, and a growth deficit. Its target proteins are disaggregated by CLPX, which also regulates heme biosynthesis via unfolding ALAS enzymes, providing access for pyridoxal-5′-phosphate (PLP). Despite efforts in diverse organisms with multiple techniques, CLPXP substrates remain controversial. Here, avoiding recombinant overexpression, we employed complexomics in mitochondria from three mouse tissues to identify endogenous targets. A CLPP absence caused the accumulation and dispersion of CLPX-VWA8 as AAA+ unfoldases, and of PLPBP. Similar changes and CLPX-VWA8 co-migration were evident for mitoribosomal central protuberance clusters, translation factors like GFM1-HARS2, the RNA granule components LRPPRC-SLIRP, and enzymes OAT-ALDH18A1. Mitochondrially translated proteins in testes showed reductions to <30% for MTCO1-3, the mis-assembly of the complex IV supercomplex, and accumulated metal-binding assembly factors COX15-SFXN4. Indeed, heavy metal levels were increased for iron, molybdenum, cobalt, and manganese. RT-qPCR showed compensatory downregulation only for Clpx mRNA; most accumulated proteins appeared transcriptionally upregulated. Immunoblots validated VWA8, MRPL38, MRPL18, GFM1, and OAT accumulation. Co-immunoprecipitation confirmed CLPX binding to MRPL38, GFM1, and OAT, so excess CLPX and PLP may affect their activity. Our data mechanistically elucidate the mitochondrial translation fidelity deficits which underlie progressive hearing impairment in PRLTS3.
Substantial progress in the field of neuroscience has been made from anaesthetized preparations. Ketamine is one of the most used drugs in electrophysiology studies, but how ketamine affects neuronal responses is poorly understood. Here, we used in vivo electrophysiology and computational modelling to study how the auditory cortex of bats responds to vocalisations under anaesthesia and in wakefulness. In wakefulness, acoustic context increases neuronal discrimination of natural sounds. Neuron models predicted that ketamine affects the contextual discrimination of sounds regardless of the type of context heard by the animals (echolocation or communication sounds). However, empirical evidence showed that the predicted effect of ketamine occurs only if the acoustic context consists of low-pitched sounds (e.g., communication calls in bats). Using the empirical data, we updated the naïve models to show that differential effects of ketamine on cortical responses can be mediated by unbalanced changes in the firing rate of feedforward inputs to cortex, and changes in the depression of thalamo-cortical synaptic receptors. Combined, our findings obtained in vivo and in silico reveal the effects and mechanisms by which ketamine affects cortical responses to vocalisations.

Background: Epilepsy surgery is an established treatment for drug-resistant focal epilepsy (DRFE) that results in seizure freedom in about 60% of patients. Correctly identifying an epileptogenic lesion in magnetic resonance imaging (MRI) is challenging but highly relevant since it improves the likelihood of being referred for presurgical diagnosis. The epileptogenic lesion’s etiology directly relates to the surgical intervention’s indication and outcome. Therefore, it is vital to correctly identify epileptogenic lesions and their etiology presurgically.
Methods: We compared the final histopathological diagnoses of all patients with DRFE undergoing epilepsy surgery at our center between 2015 and 2021 with their MRI diagnoses before and after presurgical diagnosis at our epilepsy center, including MRI evaluations by expert epilepsy neuroradiologists. Additionally, we analyzed the outcome of different subgroups.
Results: This study included 132 patients. The discordance between histopathology and MRI diagnoses significantly decreased from 61.3% for non-expert MRI evaluations (NEMRIs) to 22.1% for epilepsy center MRI evaluations (ECMRIs; p < 0.0001). The MRI-sensitivity improved significantly from 68.6% for NEMRIs to 97.7% for ECMRIs (p < 0.0001). Identifying focal cortical dysplasia (FCD) and amygdala dysplasia was the most challenging for both subgroups. 65.5% of patients with negative NEMRI were seizure-free 12 months postoperatively, no patient with negative ECMRI achieved seizure-freedom. The mean duration of epilepsy until surgical intervention was 13.6 years in patients with an initial negative NEMRI and 9.5 years in patients with a recognized lesion in NEMRI.
Conclusions: This study provides evidence that for patients with DRFE—especially those with initial negative findings in a non-expert MRI—an early consultation at an epilepsy center, including an ECMRI, is important for identifying candidates for epilepsy surgery. NEMRI-negative findings preoperatively do not preclude seizure freedom postoperatively. Therefore, patients with DRFE that remain MRI-negative after initial NEMRI should be referred to an epilepsy center for presurgical evaluation. Nonreferral based on NEMRI negativity may harm such patients and delay surgical intervention. However, ECMRI-negative patients have a reduced chance of becoming seizure-free after epilepsy surgery. Further improvements in MRI technique and evaluation are needed and should be directed towards improving sensitivity for FCDs and amygdala dysplasias.
Hyperparasitic fungi on black mildews (Meliolales, Ascomycota) : hidden diversity in the tropics

Meliolales (Sordariomycetes, Ascomycota) is a group of obligate plant parasitic microfungi mainly distributed in the tropics and subtropics. Meliolalean fungi are commonly known as “black mildews”, as they form black, superficial hyphae on the surface of vegetative and reproductive organs of vascular plants. They are considered biotrophic parasites, and the infections caused by black mildews can lead to a decrease in the photosynthetic activity of plants, as well as to an increase in the temperature and respiration rate of their leaves.
Meliolales are frequently parasitized by hyperparasitic fungi, i.e., parasitic fungi that have parasitic hosts. These hyperparasites are all Ascomycota and belong mainly to the Dothideomycetes and Sordariomycetes. Although hyperparasites represent a megadiverse group, species were only described by morphology until 1980, and the systematic position of more than 60 % of known species is still unclear. In addition, there are no DNA reference sequences available in public databases for any of the species of hyperparasites of Meliolales, and no ecological studies have been done up to now.
Before this study, no exact number of hyperparasitic fungi growing on colonies of black mildews existed. Here, we present a checklist including 189 species of fungi known to be hyperparasitic on Meliolales, but the number of existing species is likely to be even higher. The elaboration of this species checklist laid the foundations for this investigation, as it helped to understand the present state of knowledge of hyperparasitic fungi on Meliolales worldwide.
For the present study, fresh specimens of leaves infected with colonies of Meliolales and hyperparasites were opportunistically collected at 32 collection sites in Western Panama and Benin, West Africa, in 2020 and 2022, respectively. In total, 100 samples of plant specimens infected with black mildews were collected, of which 58 samples were parasitized by hyperparasitic fungi. 31 species and morphospecies of hyperparasitic fungi were identified. In addition, 35 historical specimens, including 12 type specimens, were examined for the present work.
DNA of hyperparasitic fungi was isolated directly from conidia, synnemata, apothecia, perithecia or pseudothecia of fresh and dried specimens. The main challenges faced by scientists in doing molecular studies of hyperparasitic fungi are related to the fact that the hyperparasitic fungi are intermingled with tissues of the meliolalean hosts and other organisms present in a given sample. This makes the isolation of DNA exclusively from the hyperparasite difficult. Moreover, hyperparasitic fungi on Meliolales are biotrophs and cannot be grown axenically. The hosts themselves are also biotrophic, further complicating DNA isolation from either partner. These factors have contributed to a lack of reference sequences in public databases. After more than 100 attempts, DNA of 20 specimens of hyperparasitic fungi, representing seven species, has been isolated in the context of the present investigation. Three partial nuclear gene regions were amplified and sequenced: nrLSU, nrSSU and nrITS. The datasets were assembled for phylogenetic analyses applying Maximum Likelihood (ML) and Bayesian inference (BI) methods. DNA sequences of hyperparasitic fungi on Meliolales were generated for the first time in the context of the present investigation.
Hyperparasitic fungi on Meliolales do not represent a single systematic group, but a polyphyletic ecological guild of fungi. Because of this huge diversity, only the systematics of species of perithecioid hyperparasites, as well as of the species of the genera Atractilina and Spiropes known to be hyperparasitic on black mildews was discussed in this thesis, as they represented the most common groups of fungi found in Benin and Panama. The results indicated, for example, the systematic position of Dimerosporiella cephalosporii and Paranectriella minuta in the Sordariomycetes and Dothideomycetes, respectively. In addition, the first record of a hyperparasitic fungus of black mildews in the Lecanoromycetes, namely Calloriopsis herpotricha, is reported here. The systematics of Atractilina parasitica and of some species of Spiropes is also discussed here.
In the context of the present investigation, four species new to science were described. They are presented with detailed descriptions, photos and scientific illustrations. Taxonomic studies of this thesis also generated seven new synonyms, nine new records for Benin, seven for Panama, one for Africa and two for mainland America, as well as the confirmation of one anamorph-teleomorph connection by molecular sequence data.
The ecology of hyperparasitic fungi on Meliolales is complex and far from being completely understood. The hypothesis of host specificity between hyperparasitic fungi, their meliolalean hosts and their plant hosts was tested for the first time, through a tritrophic network analysis. Results indicate that hyperparasites of Meliolales are generalists concerning genera of Meliolales, but apparently specialists at the level of order. In addition, hyperparasitic fungi tend to be found alongside their meliolalean hosts, suggesting a pantropical distribution.
Hintergrund: Bei steigender Lebenserwartung ergibt sich in Zukunft eine steigende Prävalenz der degenerativen Aortenklappenstenose (AS). Die Transkathether-Aortenklappenimplantation (TAVI) erfährt immer größere Bedeutung in der Behandlung der symptomatischen, hochgradigen AS. Eine paravalvuläre Leckage (PVL) ist ein anerkannter Risikofaktor für eine erhöhte Mortalität und sollte periinterventionell adäquat erkannt und behandelt werden. Eine Postdilatation als typische korrigierende Intervention (CI) kann allerdings zu schwerwiegenden Komplikationen führen. Weitere Instrumente zur Entscheidung über die Notwendigkeit einer CI sind erstrebenswert. Für die Aortenregurgitationsindex-Ratio (ARI-Ratio) und für die präprozedurale Kalklast, gemessen mit der Mehrschicht-Computertomografie (MSCT), wurde gezeigt, dass beide prädiktiv sind für die Notwendigkeit einer periinterventionellen CI.
Ziele: Die ARI-Ratio und der Kalkstatus wurde in Hinblick auf ihren prädiktiven Wert für die Notwendigkeit einer CI miteinander verglichen.
Methoden: Von 199 Patienten nach TAVI erhielten 38,9 % eine Portico™, 29,5 % eine Symetis Acurate™, 21,5 % eine Sapien 3™ und 10,1 % eine Evolut™. Es wurde retrospektiv der postinterventionell erhobene systolische (SB), diastolische (DB) und linksventrikuläre enddiastolische Blutdruck (LVEDP) im ARI zusammengefasst: [(DBP - LVEDP) / SBP] × 100. Die ARI-Ratio wurde berechnet als Quotient zwischen ARI nach und vor dem Eingriff. Des Weiteren wurden die MSCTs mittels „3mensio Structural Heart“ analysiert insbesondere in Hinblick auf die Verkalkung des linksventrikulären Ausflusstrakts, der Aortenklappe (gesamt sowie der einzelnen Taschen) und des sinotubulären Übergang. Dabei wurde der Kalzium-Volumen-Score auf Basis drei verschiedener Thresholds benutzt (500 Hounsfield Einheiten (HU), 800 HU und visuell-individuell). Im Folgenden wurden ROC-Kurven für den ARI-Ratio und für die verschiedenen Kalklastparameter erstellt um die Notwendigkeit einer CI zu beurteilen. Schließlich wurde die Area under the curve (AUC) der ARI-Ratio mit denen der Kalklastsurrogatparameter verglichen.
Ergebnisse: Die ARI-Ratio zeigte mit einer AUC von 0,747 das beste Ergebnis. Bei den Verkalkungsparametern zeigte die Verkalkung der noncoronaren Aortenklappentasche die beste AUC, nachfolgend die der gesamten Aortenklappe. Der 800-HU-Threshold zeigte bessere AUCs als 500 HU. Von 19 Verkalkungssurrogatparametern zeigten elf einen statistisch signifikanten Vorhersagewert auf. Die ARI-Ratio zeigte eine signifikante Überlegenheit gegenüber sechs dieser Parameter. Für die fünf Verbliebenen konnte kein signifikanter Unterschied nachgewiesen werden.
Schlussfolgerung: Die ARI-Ratio ist ein besserer Prädiktor für die Notwendigkeit einer CI als die Kalzifikationsparameter. Auch diverse Kalzifikationsparameter haben diesbezüglich eine Aussagekraft. Die Kalzifikationsparameter können bereits präinterventionell beurteilt werden können, die ARI-Ratio nur periinterventionell. Es sollte neben der Anschauung von bereits etablierten Methoden ein integrativer Ansatz gewählt werden, der Kalklast und ARI-Ratio für die Entscheidung zur Notwendigkeit einer CI miteinschließt. Weitere Studien zur Standardisierung der Verkalkungsparameter sind notwendig für eine weitere Einordnung deren Vorhersagekraft. Es bestanden teils erhebliche Unterschiede zwischen den verwendeten Klappenprothesentypen. Weitere Studien mit einer größeren Anzahl an verschiedenen Prothesentypen könnten richtungsweisende Subgruppenanalysen möglich machen.
