Grčke i semitske riječi u latinskome tekstu Ivanova evanđelja izražavaju specifične biblijske pojmove, gotovo potpuno nepoznate većini engleskog stanovništva do evangelizacije tijekom sedmoga stoljeća. Cilj je ovoga istraživanja ispitati na koje su sve načine one prevedene na staroengleski i srednjoengleski jezik te objasniti razloge takva prevođenja ondje gdje je to moguće. Komparativnom metodom istražit ćemo utjecaj porijekla riječi na njihov oblik i značenje i pokušati rasvijetliti kognitivne razlike u doživljaju biblijskih pojmova među autorima prijevoda i njihovih suvremenika, do kojih je došlo uslijed relativno dugoga razdoblja proteklog između triju prijevoda.
Die Geometrie und ihre Sprache im 16. Jahrhundert am Beispiel der deutschen volkssprachlichen Werke

Der mathematischen Wissenschaft Geometrie kam schon in der Antike eine wichtige Stelle zuteil. Die Pythagoreer (6. Jahrhundert v. Chr.) erklärten sie zu einer der vier grundlegenden Disziplinen: Arithmetik, Astronomie, Harmonielehre (Musik) und Geometrie (mathemata). Grammatik, Rhetorik, Dialektik und die vier genannten Disziplinen galten als der weltliche Wissensstoff und wurden im Mittelalter als die sieben Freien Künste (Septem artes liberales) bezeichnet und an den ersten Universitäten gelehrt. (Vgl. Taegert 2008: 13−14). "Wissenschaftssprache war im Mittelalter Latein, so dass Mathematik in lateinischer Sprache gelehrt und notiert wurde. Die Beschäftigung mit ihr war naturgemäß auf einen sehr eng umgrenzten Personenkreis beschränkt. Alltagsrelevante praktische Anwendungen der Mathematik standen hier, wie beispielweise bei der sogenannten "Klostermathematik", nicht im Vordergrund, vielmehr wurde diese Wissenschaft als eine den Geist und das schließende Denken schulende Tätigkeit angesehen." (Schuppener 2003: 40)...
Nakon uvodnih napomena o gornjomeđimurskome naselju Dragoslavec Breg (s osobitim osvrtom na povijest njegova imena, neslužbeni etnik i kretanje broja stanovnika), u radu se na osnovi terenskim i arhivskim istraživanjem prikupljene građe analizira antroponimijski sustav toga naselja. Pokazuju se značajke osobnih imena (njihova službena i neslužbena uporaba te učestalost), prezimena njegovih stanovnika (učestalost, podrijetlo, značenje i tvorbena struktura) kao i njihovi obiteljski nadimci (motivacija i tvorbene značajke).
Im vorliegenden Aufsatz werden vor dem Hintergrund der bisherigen germanistischen und bohemistischen Phraseologieforschung und in Anlehnung an die Herangehensweise von PIIRAINEN (2001) bzw. DOBROVOĽSKIJ/PIIRAINEN (2009) einige Phraseme benannt, die eine Art verstärktes Potenzial für eine geschlechtsbedingte Restriktion aufweisen. Die Überlegungen zu der Auswahl dieser Phraseme, ihrer phraseographischen Verarbeitung, der Ermittlung erforderlicher Angaben zur Geschlechtsmarkierung und einem eventuellen interlingualen phraseologischen Vergleich werden an verbalen Phrasemen mit Substantivkomponenten aus dem semantischen Bereich der Somatismen illustriert.
Die bisherige mangelhafte Berücksichtigung der substantivierten Infinitive in zweisprachigen Wörterbüchern Deutsch-Tschechisch kontrastiert mit deren oft hoher Vorkommenshäufigkeit sowie mit den Anforderungen, die an moderne Übersetzungswörterbücher seitens ihrer Benutzer gestellt werden, u. a. auch im Bereich der Darstellung der Kollokabilität und Erfassung der Synonymie bzw. Wortbildungskonkurrenz. Die Aufnahme und Darstellung der Infinitivkonvertate im entstehenden Großen Akademischen Wörterbuch Deutsch-Tschechisch wird in diesem Beitrag aus einer Corpus-Driven-Position behandelt.
During the last two decades of the 20th century writers gradually returned to traditional narrative methods, though their works were creatively enriched by the incorporation of new tendencies. The results of these efforts show traces of literary postmodernism. One of the representatives of German-language postmodern literature is Sten Nadolny, whose oeuvre is typified by its openness to the plurality and diversity of the world. His novel "Ein Gott der Frechheit" revives ancient gods and mythological figures, revealing the mysteries of old legends; this is viewed as a background to the self-destructive disorientation of contemporary society.
Wie ersichtlich gehört Saiko zu jener Schriftstellergeneration der Zwischenkriegszeit, deren Jugendjahre in die Zeit vor dem Ersten Weltkrieg fallen. Mit seiner Romandarstellung unterscheidet er sich trotz der prinzipiellen Ähnlichkeiten in wichtigen Punkten von seinen Zeitgenossen. Diese Details werden von Klaus Kastberger in seinem Artikel Einsam auf dem Floß. Wohin treibt George Saiko (Kastberger 2003: 9) in folgenderweise hervorgehoben: Obwohl die Autoren Robert Musil, Franz Kafka und James Joyce oftmals Bezugsautoren für George Saiko bildeten, grenzt Saiko sich in den wichtigsten Einzelheiten von ihnen ab, denn wie Kastberger auch erwähnt, wird alles, was in die Figuren 'hineingepackt' ist, im Laufe der Darstellung erzählerisch 'ausgepackt'. Treffender lässt es sich sagen, George Saiko hätte nicht wie Musil, Kafka und Joyce geschrieben, sondern er versucht in seinem Roman Auf dem Floß (Saiko 1980), ein Seelenpanorama der Hauptfigur zu zeichnen. Mit dem Leitgedanken der Freudschen Theorien schildert Saiko eine andere Realität und zeichnet sie in seinen Romanen nach. Das Aufschließen der menschlichen Seele, die Gründe und Abgründe des menschlichen Verhaltens werden anhand von Saikos Romancharakteren ans Licht gebracht. In seinem Roman wird der Zerfall anhand menschlicher Triebe dargestellt.
Der Aufbau des zweiten Romans der Braunschweiger Trilogie "Stopfkuchen" gründet sich auf eine Zweipoligkeit, die durch das Gegenüberstellen des Romanhelden Heinrich Schaumann und seines Gegenspielers Eduard entsteht. Diese zwei Figuren gehören gegensätzlichen Welten an. Dem Antagonisten wird nur ein Name gegeben, und zwar der Vorname: Eduard! Der Protagonist wird mit beiden Namen vorgestellt: Heinrich Schaumann! Der Name des Helden hat symbolische Bedeutung, wie man es oft bei Raabe findet: Schaumann ist der Mann, der schaut. So wird dieses Kennzeichen seiner Funktion gerecht. Der Held beobachtet etwas. Seine Beobachtungen gelten der Gesellschaft. Er beobachtet eine Gesellschaft, die er nicht gutheißt, in die er sich nicht integriert. Vom geographischen Standpunkt aus betrachtet: Der Held Schaumann ist einer der Einwohner des am Fuße des Berges Rote Schanze gelegenen Ortes namens Maiholzen.
[W]elche Rolle spielen […] Mythen in der Literatur? D.h. welche Funktionen sind mit der Rezeption von Mythen verknüpft? Diese allgemeine und einfache Fragestellung wird im Rahmen der vorliegenden Arbeit dahingehend präzisiert, dass die Bedeutungsgehalte der Mythenkomplexe in den beiden Nachkriegsromanen 'Hamlet oder Die lange Nacht nimmt ein Ende' (1946) von Alfred Döblin und 'Nekyia. Bericht eines Überlebenden' (1947) von Hans Erich Nossack auf ihre Affinität zum Geschlechteraspekt hin befragt werden. Dieser Ansatz bietet sich ganz besonders an, weil Mythen als Teil des kulturellen Gedächtnisses und somit als kulturelle Repräsentationsformen überhaupt Geschlechterverhältnisse sowie Geschlechterrollen mitprägen und demzufolge bei der Erzeugung der Geschlechterdifferenz mitbestimmend werden. Diese beiden Romane behandeln zunächst zeitgeschichtliche Problemfelder, d.h. sie setzen sich primär mit der Kriegs- bzw. Schuldfrage auseinander. Allerdings erfährt diese Fragestellung eine tiefgreifende Transformation, denn bemerkenswerterweise werden hier mithilfe der Rezeption von bestimmten Mythenkomplexen Geschlechterdiskurse, genauer Weiblichkeitsdiskurse zum Tragen gebracht, die der anfänglichen Thematik von Krieg und Schuld einen gänzlich anderen Akzent verleihen.
Im Folgenden soll es zunächst darum gehen, den Mythos aus kulturwissenschaftlicher sowie mythostheoretischer Perspektive einzuordnen. Anschließend werde ich mich auf die Analyse der genannten Romane konzentrieren.
Bu çalışmada Almanca eğitim veren ‘bölümlerimizin ihtiyacını karşılayacak nitelikteki ders materyallerinin tespiti veya yazımı için dernek çatısı altında neler yapılabileceği’ konusu irdelenmekte ve proje önerisinde bulunulmaktadır. Gerek öğrenciler gerekse öğretim elemanları nitelikli ders materyallerine gereksinim duymaktadırlar, ancak doğru ve etkili ders materyallerini elde edip uygulamak konusunda bazen sorunlar yaşanmaktadır. Bu sorunların çözümüne yönelik olarak aşağıdaki üç farklı soru doğrultusunda somut önerilerde bulunulmaktadır:
1. Bölümlerimizin gereksinim duyduğu ders materyallerinin saptanması için neler yapılabilir?
2. Bölümlerimizin gereksinim duyduğu ders materyallerinin üretilmesi için neler yapılabilir?
3. Bölümlerde özellikle okuma ve yazma becerilerine yönelik okutulan derslerde kullanılacak materyallerin üretilmesi için neler yapılabilir?
Almanca eğitim veren bölümlerin programlarında yer alan derslerde öğretim hedeflerine en uygun ders materyallerinin, alanda yetişmiş ve uzman Türk akademisyenler tarafından geliştirilmesi gerekmektedir. Üretken olmak hedefinin bir gereği olarak dernek üyelerinden görüş toplayıp, eksiklikleri belirleyip, derslerimize uygun materyal üretimi projesini gerçekleştirmek üniversitelerimizin Almanca bölümlerinde görev yapan dernek üyelerimizin somut katkılarıyla oluşabilecektir.
Gardening is a popular pastime, but commercial horticulture is responsible for the introduction of alien species and contributes to invasions in a variety of ways. Although an extensive international literature is available on plant invasions, it is still important at the national level to examine the influence of local factors. Accordingly, 17 nurseries in Estonia that cultivated and sold perennial alien species were selected, and a list of species and prices was compiled. The relationships between species status, and factors such as their abundance in the wild were examined statistically. A qualitative list of the nationally problematic species among herbaceous perennials was also completed. A total of 880 taxa were recorded, of which 10.3% were native and 89.7% alien. In all, 87.3% of the alien species were still confined to cultivated areas. The ecological and socio-economic characteristics of the taxa were described, and lists of the families of casual, naturalised and invasive aliens were provided. Both native and increasing wild alien species have a very similar profile on the market. Alien species that are less expensive, widely available and have more cultivars per species on the market are also more likely to escape. The invasive status and abundance of escaped aliens in an area increases with residence time. In general, socio-economic factors create new and reflect previous propagule pressures from commercial horticulture, which continuously increase the likelihood of alien species surviving and invading new areas. Our findings suggest that these national socioeconomic market-related factors explain much of the invasiveness of various perennial ornamental species, and therefore regional and national authorities urgently need to regulate and control the ornamental plant trade to diminish the risk of new invasions.
Türk üniversitelerindeki Alman dili ve edebiyatı bölümleri bir süredir kriz içinde. Zira ülkemizde Alman dili ve edebiyatı bölümü mezunlarının iş arama ve bulmada yaşadıkları maddi ve manevi sıkıntılar, üniversiteye girme aşamasındaki Almanca bilen gençlerimizin bu bölümlere olan ilgilerinin giderek azalmasına, mezun olduktan sonra kendilerine daha iyi olanak sunacak farklı bölümlere girmeyi tercih etmelerine neden olmaktadır. Ülkemizde üniversiteye girecek olan öğrencilerin Alman dili ve edebiyatı bölümlerinden aldıkları diplomaları ile ulusal ve uluslararası alanlarda arzu edilen şartlarda iş bulabilmeleri için Alman dili ve edebiyatı bölümlerinin programları yeniden gözden geçirilmektedir. Bu yazıda da Alman dili ve edebiyatı bölümlerinin konumu, bu bölümlerde okuyan öğrencilerimizin sorunları, nedenleri ve bu sorunların üstesinden gelmek için önerilen düşünceler tartışılacaktır.

3) XII. Uluslararası Türk Germanistik Kongresinin planlanması gündemli oturumlarda tartışılan ve benimsenen görüşler aşağıda sunulmuştur.
In 'Mein Jenseits', der dritten Novelle innerhalb Walsers umfangreicher literarischer Produktion, werden uns von Walsers früheren Büchern bekannte Themen aus einer neuen, einem "Spätwerk" entsprechenden Perspektive dargeboten, wie im Laufe dieser Analyse gezeigt werden soll. In diesem Zusammenhang soll besonders die zentrale Rolle, die hier einem Bild für das Textganze zukommt, beleuchtet werden.
ω(782) und ϕ(1020) Mesonenproduktion durch Dielektronen in pp-Kollisionen bei √s = 7 TeV mit ALICE
(2013)
Die Niedrigmassendielektronen (Elektron-Positron Paare mit kleiner invarianten Masse) sind wichtige experimentelle Sonden, um die Eigenschaften des in ultra-relativistischen Schwerionenkollisionen erzeugten heißen und dichten Mediums zu untersuchen. Elektronen koppeln nicht an die starke Wechselwirkung, weshalb sie wichtige Informationen über die gesamten Kollisionsphasen geben. Die Zerfälle von ω(782) und ϕ(1020)-Mesonen in Dielektronen ermöglichen es, besonders wichtige Informationen über ihre In-Medium-Eigenschaften zu erhalten, da Proton-Proton (pp)-Kollisionen als mediumfreie Referenz angenommen werden. Außerdem sind pp-Kollisionen auch für sich genommen interessant, um die Teilchenproduktion im Energiebereich des LHC (Large Hadron Collider) zu untersuchen.
In dieser Analyse werden die Elektronen im mittleren Rapiditätsbereich von |η| < 0.8 mit ITS (Inner Tracking System), TPC (Time Projection Chamber) und TOF (Time of Flight) gemessen.
Die transversalen Impulsspektren der ω(782) und ϕ(1020)-Mesonen im e+e--Zerfallskanal in pp-Kollisionen bei p √s = 7 TeV werden gezeigt. Das transversale Impulsspektrum des ω(782)-Mesons im e+e--Zerfallskanal wird mit den pT-Spektren in den µ+µ--und in den π0π+π--Zerfallskanälen verglichen, während das pT-Spektrum vom ϕ(1020)-Meson im e+e--Zerfallskanal mit den pT-Spektren in µ+µ-- und K+K--Zerfallskanälen verglichen wird.
Çeviri yaparken her iki dile, kültüre yeterince hâkim olmamak bir dizi çeviri hatalarına yol açabilir. Bu noktada kaynak ve erek dilde yeterli kelime hazinesine sahip olmanın yanı sıra kelimelerin kullanım alanlarını, kurallarını, edim bilimsel etkilerini, sözdizimsel kuralları vs. de iyi bilmek gerekir. Humboldt ve Saussure’ün dil hakkındaki düşüncelerinden etkilenen Trier’in ortaya attığı sözlüksel alan teorisinin metin anlama ve anlatma edinci kapsamında etkilerinin neler olabileceği ve çeviribilimin sözlüksel alan teorisinden nasıl yararlanabileceği konusu irdelenmeye çalışılacaktır. Trier’e (1973:5) göre bir sözcüğün anlaşılabilmesi için, sözlüksel alanın tamamının bilinmesi
gerekir ve ancak sözlüksel alana hâkim isek o sözcüğü doğru anlayabiliriz. Anlam sadece ve sadece sözlüksel alan sayesinde vardır. Sözlüksel alan yoksa anlam da yoktur. Anlatılmak istenen düşünceye veya olguya dair bir kelimenin belli bir dilde bulunmaması bu düşüncenin veya olgunun o dilde olmadığı anlamına gelmez. Hayata dair genel kültür bilgimize ve tecrübelerimize dayanarak bu yeni kavramı
anlayabiliriz.
Wie bekannt, besitzt neben der literarischen Übersetzung auch die fachsprachliche Übertragung einen wichtigen Platz in der übersetzungswissenschaftlichen Forschung. In Rahmen des Themas Übersetzung und Bearbeitung soll in diesem Beitrag festgestellt werden, welche satzstrukturelle Abwandlungen das neue Strafgesetzbuch der Türkei (YTCK) vollzogen hat.

Gerçek yaşamla ilgili bu saptama, çağdaş Alman yazarlarından Karl-Heinz Ott'un 1998 yılında yayımlanmış ve "Ins Offene" adını taşıyan romanının kurmaca dünyasında, bu sefer anne-oğul ilişkisi bazında, tersine işlemektedir.
Çalışmada, anne ve babanın birlikte yaşamamaları, yani aralarında evliliğin gerçekleşmemesi, anne ve oğlunun farklı karakterde olmaları, annenin oğluna olan aşırı bağımlılığı ve kırsal ataerkil toplum bu ilişkiyi sorunlu hale getiren ana etmenler olarak belirlenmiştir.
Ausdrucksphänomene fluktuieren zwischen den Sphären von Körperlichkeit und Bewegung sowie Sprachlichkeit und Kommunikation. Um Ausdruck in seiner eigentümlichen Mobilität und Wandlungsfähigkeit zu erfassen, erkunden die Beiträge des vorliegenden Bands seine ästhetischen, sozialen und (inter-)subjektiven Dimensionen und deren Überschneidungen sowohl im Rahmen historischer Entwicklungen als auch vor dem Hintergrund aktueller Erkenntnisse und Debatten der Psychologie, Philosophie, Hirnforschung und Linguistik. Aus den Themenbereichen Körper, Sprache und Künste, aber gleichfalls aus übergreifenden Fragestellungen der Anthropologie heraus werden kommunikative und körperliche Ausdrucksformen, auch in ihrem Zusammenspiel, in systematischer Weise analysiert und auf ihre situative und historische Spezifik hin untersucht. Nicht nur sprachliche Äußerungen, sondern ebenso Gesten und Gebärden, Tanz, Musik, Mienenspiel und Malerei stoßen dabei an Grenzen der Verständlichkeit und verweisen auf Ausdrucksqualitäten jenseits von Verbalität und Körperlichkeit.
The aim of this study is to examine the different grades students’ understanding levels of the concept of religion in elementary education. A total of 107 different grades students taken from elementary schools were asked the concept in using open ended question developed by the researcher. Obtained data showed that students couldn’t understand the concept correctly and scientifically and the majority of the students had a misconception about the concept such as worshipping, worships, being ethical, ethical behaviours and obligatory behaviours. Furthermore, some students had specific conceptual confusions about the concept.
Spread across a number of countries around the world, and concentrated in four Middle Eastern countries, Kurds have yearned for their own country for almost a century, but were forgotten when the region was carved up by the Sykes-Picot Agreement early in the twentieth century. Since then, the creation of a Kurdish state was high on the agenda of all Kurds. This was especially true when we consider the lot of Kurds in Turkey, Syria, Iraq and Iran. This book examines the political situation of Kurds in these four countries, looks at how this has changed-particularly in the past decade-and considers what the future might hold for the Kurdish people and for the notion of a state of Kurdistan. It asks the question of whether a Kurdish state is achievable, or, even, desirable. The book is written for policy makers and academics interested in the Middle East region and in Kurdish politics in particular. It is written in an accessible way that makes it easy reading for anyone curious about the region and its people.

Im Gegensatz zu den anderen Wirbeltieren spielt sich das Leben der Fische ausschließlich im Wasser ab und bleibt den Blicken des Menschen verborgen. Systematische Arterfassungen werden erst seit jüngerer Zeit durchgeführt, aber jede Methode der Bestandserfassung liefert nur ein unvollkommenes Bild. Daher blieb es auch lange Zeit unbekannt, dass etliche Fischarten nur noch in geringen Restbeständen vorkommen, einige sogar völlig verschwunden sind. Als diese Situation bekannt wurde (Paepke 1981), erregte sie kaum Aufmerksamkeit. Erst in den 1980er Jahren begannen auch im heutigen Sachsen-Anhalt Fischkundler mit der systematischen Erfassung der Vorkommen von Fischen und Rundmäulern (Engelke 1988, Wüstemann 1989, Zuppke 1986). Nach 1990 wurden diese Bemühungen dank der Verfügbarkeit effektiverer Fanggeräte intensiviert.
Im soeben erschienenen Handbuch der FFH-Gebiete Sachsen-Anhalts (Jentzsch & Reichhoff 2013) wird für das FFH-Gebiet "Feuchtwiese bei Dobien" (FFH0250LSA) angegeben, dass keine Kenntnisse zur Fauna vorliegen. Daher erscheint es angebracht, über das dortige Vorkommen einer gefährdeten Tagfalterart zu informieren. Aim: The aim of this study was to measure cortico-cortical connectivity in multiple sclerosis (MS) patients by TMS-evoked potential (TEP) latencies in EEG evoked by transcranial magnetic stimulation (TMS) of the hand area of the primary motor cortex of one hemisphere. TEPs were recorded on the stimulated- and at the homologue site in the non-stimulated contralateral hemisphere. Both interhemispheric directions were tested. Interhemispheric latencies of the two main reproducible TEPs, the positive component at 60 ms and the negative component at 100 ms (P60 and N100, respectively), were expected to be significantly prolonged in MS-patients compared to healthy volunteers.
Material and methods: The study compared interhemispheric propagation of P60 and N100 in groups of 12 patients with early-stage relapsing-remitting MS (RRMS) and 16 age- and gender-matched healthy controls. The study was approved by the Ethics Committee of the Medical Faculty of the Goethe-University of Frankfurt/Main and conformed to the latest revision of the Declaration of Helsinki of 2008. TEPs were recorded by means of EEG and their latencies were statistically evaluated in 10 channels around the stimulation site and in 10 corresponding electrodes in the non-stimulated contralateral hemisphere. Interhemispheric conduction time was calculated by the difference of TEP latency in non-stimulated vs. stimulated hemisphere.
Results: An ANOVA on interhemispheric conduction time showed a significant prolongation for the N100 from left to right hemisphere in MS compared to controls, while no group differences were found for the P60 and the N100 from right to left hemisphere.
Conclusion: The results provide first evidence that the N100 may constitute an interesting marker to measure interhemispheric conduction delays in early-stage RRMS. The specificity of the present finding and its relation to fiber tract pathology should be examined in further correlative analyses with diffusion tensor imaging and other structural MRI data.
Background: Exercise seems to minimize prostate cancer specific mortality risk and treatment related side effects like fatigue and incontinence. However the influence of physical activity on the immunological level remains uncertain. Even prostate cancer patients undergoing palliative treatment often have a relatively long life span compared to other cancer entities. To optimize exercise programs and their outcomes it is essential to investigate the underlying mechanisms. Further, it is important to discriminate between different exercise protocols and therapy regimes.
Methods/Design: The ProImmun study is a prospective multicenter patient preference randomized controlled trial investigating the influence of a 24 week endurance exercise program in 80–100 prostate cancer patients by comparing patients undergoing Antiandrogen therapy combined with exercise (AE), Antiandrogen therapy without exercise (A), Chemotherapy with exercise(CE) or Chemotherapy without exercise (C). The primary outcome of the study is a change in prostate cancer relevant cytokines and hormones (IL-6, MIF, IGF-1, Testosterone). Secondary endpoints are immune cell ratios, oxidative stress and antioxidative capacity levels, VO2 peak, fatigue and quality of life. Patients of the intervention group exercise five times per week, while two sessions are supervised. During the supervised sessions patients (AE and CE) exercise for 33 minutes on a bicycle ergometer at 70-75% of their VO2 peak. To assess long term effects and sustainability of the intervention two follow-up assessments are arranged 12 and 18 month after the intervention.
Discussion: The ProImmun study is the first trial which primarily investigates immunological effects of a six month endurance exercise program in prostate cancer patients during palliative care. Separating patients treated with Antiandrogen therapy from those who are additionally treated with Chemotherapy might allow a more specific view on the influence of endurance training interventions and the impact of different therapy protocols on the immune function.

Unter ökonomischen Gesichtspunkten ist die Mainmetropole ein etablierter Global Player, nun will sie auch in Sachen Klimaschutz eine Vorreiterrolle einnehmen. Ziel ist es, die städtische Energieversorgung in den nächsten vier Jahrzehnten komplett aus erneuerbaren Ressourcen sicherzustellen. Dabei ist Frankfurt bereits jetzt auf einem guten Weg. Neben Kopenhagen und Bristol schaffte es die größte Stadt Hessens in die Finalrunde um den Titel „Europäische Grüne Hauptstadt 2014“.
Metallorganische Netzwerke (engl. metal-organic frameworks, MOFs) sind eine neuartige Klasse mikro/mesoporöser Materialien, für die eine Vielzahl von möglichen Anwendungen demonstriert werden konnte. Das Ziel dieser Arbeit besteht in der Synthese von MOF Mikro/Nanopartikeln sowie der Herstellung von sogenannten Oberflächen-deponierten MOFs (engl. surface-attached metal-organic frameworks, SURMOFs). MOF Partikel mit kontrollierbarer Morphologie und Größe wurden unter milden Bedingungen synthetisiert. Um MOFs als Sensoren, intelligente Membrane, oder in nanotechnologischen Bauelementen verwenden zu können, ist die Integration auf der jeweiligen Oberfläche wichtig. Daher beschäftigt sich der Großteil dieser Arbeit mit der kontrollierten Abscheidung von SURMOFs auf verschiedenartigen Trägermaterialien. Etliche interessante Eigenschaften (z.B. die Fluoreszenz in Abhängigkeit von der Gegenwart von Gastmolekülen und die dynamische Gasadsorptionskapazität) der SURMOFs wurden untersucht.
The information processing abilities of neural circuits arise from their synaptic connection patterns. Understanding the laws governing these connectivity patterns is essential for understanding brain function. The overall distribution of synaptic strengths of local excitatory connections in cortex and hippocampus is long-tailed, exhibiting a small number of synaptic connections of very large efficacy. At the same time, new synaptic connections are constantly being created and individual synaptic connection strengths show substantial fluctuations across time. It remains unclear through what mechanisms these properties of neural circuits arise and how they contribute to learning and memory. In this study we show that fundamental characteristics of excitatory synaptic connections in cortex and hippocampus can be explained as a consequence of self-organization in a recurrent network combining spike-timing-dependent plasticity (STDP), structural plasticity and different forms of homeostatic plasticity. In the network, associative synaptic plasticity in the form of STDP induces a rich-get-richer dynamics among synapses, while homeostatic mechanisms induce competition. Under distinctly different initial conditions, the ensuing self-organization produces long-tailed synaptic strength distributions matching experimental findings. We show that this self-organization can take place with a purely additive STDP mechanism and that multiplicative weight dynamics emerge as a consequence of network interactions. The observed patterns of fluctuation of synaptic strengths, including elimination and generation of synaptic connections and long-term persistence of strong connections, are consistent with the dynamics of dendritic spines found in rat hippocampus. Beyond this, the model predicts an approximately power-law scaling of the lifetimes of newly established synaptic connection strengths during development. Our results suggest that the combined action of multiple forms of neuronal plasticity plays an essential role in the formation and maintenance of cortical circuits.

Significant progress has been made over the last decade towards realizing the potential of natural killer (NK) cells for cancer immunotherapy. NK cells can respond rapidly to transformed and stressed cells, and have the intrinsic potential to extravasate and reach their targets in almost all body tissues. In addition to donor-derived primary NK cells, also continuously expanding cytotoxic cell lines such as NK-92 are being considered for adoptive cancer immunotherapy. High cytotoxicity of NK-92 has previously been shown against malignant cells of hematologic origin in preclinical studies, and general safety of infusion of NK-92 cells has been established in phase I clinical trials. To enhance their therapeutic utility, we genetically modified NK-92 cells to express chimeric antigen receptors (CAR) specific for tumor-associated surface antigens. Such CAR were composed of a tumor-specific scFv antibody fragment fused via hinge and transmembrane domains to intracellular signaling moieties such as CD3 zeta chain, or composite fusion molecules also containing a costimulatory protein domain in addition to CD3 zeta. For development towards clinical applications, here a codon-optimized second generation CAR was constructed that consists of an ErbB2-specific scFv antibody domain fused via a linker to a composite CD28-CD3 zeta signaling domain. GMP-compliant protocols for vector production, lentiviral transduction and expansion of a genetically modified NK-92 single cell clone (NK-92/5.28.z) were established. Functional analysis of NK-92/5.28.z cells revealed high and stable CAR expression, selective cytotoxicity against ErbB2-expressing but otherwise NK-resistant tumor cells of different origins in vitro, as well as homing to ErbB2-expressing tumors in vivo. Furthermore, antigen specificity and selective cytotoxicity of these cells were retained in vivo, resulting in antitumoral activity against subcutaneous and intracranial glioblastoma xenografts in NSG mice. Ongoing work now focuses on the development of these cells for adoptive immunotherapy of ErbB2-positive glioblastoma.
This work presents a biochemical, functional and structural characterization of Aquifex aeolicus F1FO ATP synthase obtained using both a native form (AAF1FO) and a heterologous form (EAF1FO) of this enzyme.
F1FO ATP synthases catalyze the synthesis of ATP from ADP and inorganic phosphate driven by ion motive forces across the membrane and therefore play a key cellular function. Because of their central role in supporting life, F1FO ATP synthases are ubiquitous and have been remarkably conserved throughout evolution. For their biological importance, F1FO ATP synthases have been extensively studied for many decades and many of them were characterized from both a functional and a structural standpoint. However, important properties of ATP synthases – specifically properties pertaining to their membrane embedded subunits – have yet to be determined and no structures are available to date for the intact enzyme complex. Therefore, F1FO ATP synthases are still a major focus of research worldwide. Our research group had previously reported an initial characterization of AAF1FO and had indicated that this enzyme presents unique features, i.e. a bent central stalk and a putatively heterodimeric peripheral stalk. Based on such a characterization, this enzyme revealed promising for structural and functional studies on ATP synthases and became the focus of this doctoral thesis. Two different lines of research were followed in this work.
First, the characterization of AAF1FO was extended by bioinformatic, biochemical and enzymatic analyses. The work on AAF1FO led to the identification of a new detergent that maintains a higher homogeneity and integrity of the complex, namely the detergent trans-4-(trans-4’-propylcyclohexyl)cyclohexyl-α-D-maltoside (α-PCC). The characterization of AAF1FO in this new detergent showed that AAF1FO is a proton-dependent, not a sodium ion-dependent ATP synthase and that its ATP hydrolysis mechanism needs to be triggered and activated by high temperatures, possibly inducing a conformational switch in subunit γ. Moreover, this approach suggested that AAF1FO may present unusual features in its membrane subunits, i.e. short N-terminal segments in subunits a and c with implications for the membrane insertion mechanism of these subunits.
Investigating on these unique features of A. aeolicus F1FO ATP synthase could not be done using A. aeolicus cells, because these require a harsh and dangerous environment for growth and they are inaccessible to genetic manipulations. Therefore, a second approach was pursued, in which an expression system was created to produce the enzyme in the heterologous host E. coli. This second approach was experimentally challenging, because A. aeolicus F1FO ATP synthase is a 500-kDa multimeric membrane enzyme with a complicated and still not entirely determined stoichiometry and because its encoding genes are scattered throughout A. aeolicus genome, rather than being organized in one single operon. However, an artificial operon suitable for expression was created in this work and led to the successful production of an active and fully assembled form of Aquifex aeolicus F1FO ATP synthase. Such artificial operon was created using a stepwise approach, in which we expressed and studied first individual subunits, then subcomplexes, and finally the entire F1FO ATP synthase complex. We confirmed experimentally that subunits b1 and b2 form a heterodimeric subcomplex in the E. coli membranes, which is a unique case among ATP synthases of non-photosynthetic organisms. Moreover, we determined that the b1b2 subcomplex is sufficient to recruit the soluble F1 subcomplex to the membranes, without requiring the presence of the other membrane subunits a and c. The latter subunits can be produced in our expression system only when the whole ATP synthase is expressed, but not in isolation nor in the context of smaller FO subcomplexes. These observations led us to propose a novel mechanism for the assembly of ATP synthases, in which first the F1 subcomplex attaches to the membrane via subunit b1b2, and then cring and subunits a assemble to complete the FO subcomplex. Furthermore, we could purify the heterologous ATP synthase (EAF1FO) to homogeneity by chromatography and electro-elution. Enzymatic assays showed that the purified form of EAF1FO is as active as AAF1FO. Peptide mass fingerprinting showed that EAF1FO is composed of the same subunits as AAF1FO and all soluble and membrane subunits could be identified. Finally, single-particle electron microscopy analysis revealed that the structure of EAF1FO is identical to that of AAF1FO. Therefore, the EAF1FO expression system serves as a reliable platform for investigating on properties of AAF1FO.
Specifically, in this work, EAF1FO was used to study the membrane insertion mechanism of rotary subunit c. Subunits c possess different lengths and levels of hydrophobicity across species and by analyzing their N-terminal variability, four phylogenetic groups of subunits c were distinguished (groups 1 to 4). As a member of group 2, the subunit c from A. aeolicus F1FO ATP synthase is characterized by an N-terminal segment that functions as a signal peptide with SRP recognition features, a unique case for bacterial F1FO ATP synthases. By accurately designing mutants of EAF1FO, we determined that such a signal peptide is strictly necessary for membrane insertion of subunit c and we concluded that A. aeolicus subunit c inserts into E. coli membranes using a different pathway than E. coli subunit c. Such a property may be common to other ATP synthases from extremophilic organisms, which all cluster in the same phylogenetic group.
In conclusion, the successful production of the fully assembled and active F1FO ATP synthase from A. aeolicus in E. coli reported in this work provides a novel genetic system to study A. aeolicus F1FO ATP synthase. To a broader extent, it will also serve in the future as a solid reference for designing strategies aimed at producing large multi-subunit complexes with complicated stoichiometry.

Hermann Hesse erzählt von "brausenden Lachsalven", die Zuschauer seien "wie Besessene vom Dämon gestoßen" worden, so sehr hätten sie gelacht. Kurt Tucholsky beschreibt einen Saal voller Lachen; Rudolf Frank berichtet von "Wogen unbändigen Lachens, die das Haus überfluteten". Und Bertolt Brecht schüttelte sich, wie er sich erinnert, vor Lachen. Im Publikum saßen begeistert die Brüder Mann, Alfred Polgar, Franz Blei, Carl Zuckmayer, Samuel Beckett (der "recht traurig viel gelacht" habe) und nicht wenige der Kulturgrößen der 1920er und 1930er Jahre. Brecht, der mit Valentin auch auf dem Oktoberfest spielte und ihm einen entscheidenden Hinweis für den Verfremdungseffekt verdankte, bezeichnete ihn als eine der "eindringlichsten Figuren der Zeit". Die Literaturgeschichten hingegen schreiben Karl Valentin keine besondere Bedeutung zu - wie sie überhaupt der Komik wenig Aufmerksamkeit widmen. Die Geringschätzung einer "Unterhaltung" verweist die komische Figur von der Bühne des Bedeutsamen: Einem Publikumserfolg mißtraut der eifersüchtige Ästhet, der sonst das Wort "Volk" politisch-moralisch gern im Munde führt.
Split intein enabled protein trans-splicing (PTS) is a powerful method for the ligation of two protein fragments, thereby paving the way for various protein modification or protein function control applications. PTS activity is strongly influenced by the amino acids directly flanking the splice junctions. However, to date no reliable prediction can be made whether or not a split intein is active in a particular foreign extein context. Here we describe SPLICEFINDER, a PCR-based method, allowing fast and easy screening for active split intein insertions in any target protein. Furthermore we demonstrate the applicability of SPLICEFINDER for segmental isotopic labeling as well as for the generation of multi-domain and enzymatically active proteins.
Background: Novel microscopic techniques which bypass the resolution limit in light microscopy are becoming routinely established today. The higher spatial resolution of super-resolution microscopy techniques demands for precise correction of drift, spectral and spatial offset of images recorded at different axial planes.
Methods: We employ a hydrophilic gel matrix for super-resolution microscopy of cellular structures. The matrix allows distributing fiducial markers in 3D, and using these for drift correction and multi-channel registration. We demonstrate single-molecule super-resolution microscopy with photoswitchable fluorophores at different axial planes. We calculate a correction matrix for each spectral channel, correct for drift, spectral and spatial offset in 3D.
Results and discussion: We demonstrate single-molecule super-resolution microscopy with photoswitchable fluorophores in a hydrophilic gel matrix. We distribute multi-color fiducial markers in the gel matrix and correct for drift and register multiple imaging channels. We perform two-color super-resolution imaging of click-labeled DNA and histone H2B in different axial planes, and demonstrate the quality of drift correction and channel registration quantitatively. This approach delivers robust microscopic data which is a prerequisite for data interpretation.
