Hintergrund: Die Interaktion zwischen β-HCG und TSH in der Schwangerschaft stellt ein differentialdiagnostisches Problem dar, weil die Wertung von supprimierten TSH-Spiegeln erschwert wird. Dies kann im schlimmsten Fall zu Fehlinterpretationen führen. Ziel der vorliegenden Arbeit war, diese Interaktion an einem großen Kollektiv in einen zeitlichen Kontext mit dem Verlauf der Schwangerschaft zu bringen, da der Zeitpunkt des Screenings entscheidenden Einfluss auf die Höhe des TSH-Spiegels hat. Zusätzlich wurden anhand der vorliegenden Daten Referenzbereiche für Schwangere berechnet und der Einfluss einer Jodmedikation untersucht.
Patienten und Methoden: Aus einem unselektionierten Pool von Patientinnen eines nuklearmedizinischen Praxisverbundes wurden die Daten von 1283 schilddrüsengesunden Schwangeren zwischen 16 und 48 Jahren ausgewertet. Neben der TSH-Bestimmung lag ein Schwerpunkt auf dem zeitlichen Verlauf, so dass die Schwangeren in Untergruppen von je 2 Wochen analysiert wurden. Untersucht wurde der Einfluss der Jodmedikation auf die TSH-Werte. Am Ende erfolgte mit Hilfe der logarithmischen Transformation unter Verwendung der 2-Sigma-Grenzen die Bestimmung neuer TSH-Referenzbereiche für Schwangere.
Ergebnisse: Es zeigt sich zu Beginn der Schwangerschaft ein Anstieg der mittleren TSH-Werte von 1,22 mU/l in der 2. SSW bis auf 1,7 mU/l um die 7. SSW mit einem konsekutiven Abfall der TSH-Werte bis auf 0,9 mU/l bis zur 16. SSW (entsprechend 52,9%). Der größte Abfall findet sich in der 12. bis 14. SSW, also zum Zeitpunkt des ersten Screenings. Die Jodmedikation hat keinen maßgeblichen Einfluss auf den TSH-Wert. Die Berechnung schwangerschaftskorrigierter Referenzbereiche zeigt im ersten Drittel TSH-Werte von 0,08 – 3,67 mU/l, im 2. Drittel 0,04 – 2,88 mU/l und im 3. Trimenon 0,17 – 3,19 mU/l.
Schlussfolgerungen: Die Arbeit zeigt, dass die niedrigsten TSH-Werte zum Zeitpunkt des ersten Screenings zu erwarten sind und deswegen möglicherweise zu Fehlentscheidungen führen können. Ein relevanter Zusammenhang der Jodmedikation mit dem TSH-Wert lässt sich nicht nachweisen. Neue Referenzbereiche für Schwangere könnten hilfreich sein, dieses diagnostische Dilemma zu vermeiden.
Habituation ist eine der einfachsten Formen des Gedächtnisses. Hierbei handelt es sich um die erlerne Gewöhnung an einen harmlosen Reiz. Dies bedeutet, dass nach mehrfacher wiederholter Repräsentation eines harmlosen Reizes die Reaktion darauf stetig abnimmt, bis sie völlig zum erliegen kommt. Je nach Trainingsprotokoll kann diese Gewöhnung bis zu mehren Tagen andauern. Habituation ist hoch konserviert und ein Verhaltensmuster, dass auch bei sehr einfachen vielzelligen Organismen zu finden ist und untersucht werden kann. Zur Untersuchung des Zusammenspiels innerhalb eines neuronalen Netzwerkes, welches für die Habituation des Rückzugsreflexes (Ausweichreaktion nach Berührung) verantwortlich ist wurde hier der Fadenwurm Caenohabditis elegans (C. elegans) als Modell Organismus verwendet. Aufgrund seines einfachen, nur 302 Zellen umfassenden, Nervensystems eignet sich C. elegans sehr gut für Grundlagenforschung in diesem Bereich. Das neuronale Netzwerk, das verantwortlich ist für den Rückzugsreflex ist in drei Ebenen organisiert. Wahrgenommen wird der Reiz von sensorischen Neuronen (ASH, ALM, AVM, PLM, PVM). Die Weiterleitung erfolgt über verschiedene Interneuronen (AVA, AVB, AD, AVE, PVC) hin zu den Motorneuronen, welche die Muskeln enervieren und somit die Reaktion auf den in erster Ebenen wahrgenommen Reiz auslösen.
Mit Hilfe von optogenetischen Werkzeugen wurde hier Untersucht welche Rolle einzelne Zellen innerhalb dieses Netzwerkes innehaben und an welcher Stelle innerhalb des Netzwerkes die kurzzeitige Habituation des Reizes, nach einem Einfachen Lernprotokoll stattfindet. Zuerst musste eine Möglichkeit gefunden werden die zur Verfügung stehenden optogenetischen Werkzeuge zellspezifisch zu exprimieren. In dieser Arbeit wurden hierfür Rekombinasesysteme verwendet, die es ermöglichten zur Expression eine Kombination aus 2 verschiedenen Promotoren zu verwenden. Beide Promotoren dürfen hierbei nur in einer Zelle, der Zielzelle, überlappen. Es konnte zellspezifische Expression des Kationenkanals Chanelrhodopsin 2 (ChR2) in den beiden Zellparen AVAL/R und ASHL/R (nimmt aversive Reize wahr) erreicht werden.
Zur Untersuchung der Habituation wurde zusätzlich noch ein Wurmstamm verwendet, welcher ChR2 unter dem mec-4 Promotor exprimiert. ChR2 ist hier in den Mechanorezeptorneuronen (MRN) ALM, AVM, PLM und PVM exprimiert. Die hier durchgeführten Experimente deuten darauf hin das den MRNs die Größte Rolle bei der Ausbildung einer Habituation zukommt. Es gibt jedoch auch Hinweise darauf, dass AVA zusätzlich eine Rolle spielt.
Im weiteren Verlauf der Arbeit wurde die Rolle von AVA genauer untersucht. AVA gilt als der Hauptsignalgeber für eine Rückwärtsbewegung (spontan und nach Reizempfang). Es konnte gezeigt werden dass eine Unterbrechung der ’Gap Junktionen’ zwischen AVA und PVC eine stärkere Reaktion zur Folge haben. AVA scheint also durch PVC inhibiert zu werden. Ebenfalls mit AVA direkt interagierende Neuronen sind AVD und AVE. Mit den hier zur Verfügung stehenden Mitteln konnte die genaue Modulation von AVA durch diese Zellen jedoch nicht gezeigt werden.
In dieser Arbeit konnte der Grundstein für eine funktionale Aufklärung des Nervensystems von C. elegans gelegt werden. Vor allem durch die Möglichkeit der zellspezifischen Expression kann es zukünftig gelingen das Zusammenspiel der einzelnen Nervenzellen und ihren Anteil an einem bestimmtem Verhalten zu Untersuchen.
Im Rahmen der hier vorliegenden Arbeit wurde der Einfluss sowohl akuter als auch chronischer Aufnahme subletaler Mengen des Insektizids Thiacloprid auf Einzelbienen und Bienenvölker untersucht. Anlass für diese Art an Untersuchungen gibt ein seit Jahren in Nordamerika und Europa auftretendes unerklärliches Phänomen, „Colony Collapse Disorder“ genannt, bei dem Bienenvölker durch einen plötzlichen Verlust der Flugbienen zusammenbrechen. Als Ursache für das Völkersterben stehen neben anderen Faktoren wie Parasiten, Pathogenen und Umweltfaktoren die Insektizide aus der Gruppe der Neonikotinoide und deren Auswirkungen auf Bienen in subletalen Mengen im Verdacht. Basierend auf Studien der Europäischen Behörde für Lebensmittelsicherheit (EFSA) wurde der zugelassene Einsatz der drei Neonikotinoide Clothianidin, Imidacloprid und Thiamethoxam im Pflanzenschutz für zunächst zwei Jahre durch die EU-Kommission stark eingeschränkt.
Thiacloprid, ein weiteres Insektizid, welches zur Gruppe der Neonikotinoide gehört, ist weiterhin für den Einsatz im Pflanzenschutz zugelassen. Es wirkt in ähnlicher Weise wie die zuvor genannten Neonikotinoide als Agonist am nikotinischen Acetylcholinrezeptor, wobei es jedoch als weniger toxisch für Bienen gilt. Trotzdem sind subletale Auswirkungen dieses Neonikotinoids auf Bienen denkbar, die sich in Verhaltensänderungen der Bienen äußern und als Folge Einfluss auf das gesamte Bienenvolk nehmen könnten.
In der hier vorliegenden Arbeit wurden in chronisch mit Thiacloprid eingefütterten Völkern über mehrere Monate regelmäßige Populationsschätzungen durchgeführt, um die Entwicklung der Bienenvölker unter Aufnahme von Thiacloprid festzustellen. In einem weiteren Versuch wurde die Entwicklung der Brut unter chronischer Fütterung mit Thiacloprid beobachtet. Zusätzlich wurde eine große Zahl an Bienen mit RFID-Transpondern ausgestattet, um das Flugverhalten zu dokumentieren. Insbesondere wurden hier der Zeitpunkt des ersten Ausflugs und die Lebensdauer der Bienen zu Vergleichen herangezogen. Nach akuter Fütterung einer subletalen Einzeldosis Thiacloprid wurden Versuche zum Heimkehrvermögen von Bienen durchgeführt.
Unter feldrelevanten und bis zu zehnfach höheren Thiacloprid-Konzentrationen wurden keine beeinträchtigenden Einflüsse auf die Volksentwicklung beobachtet. Bei Konzentrationen, die um ein 25faches bzw. ein 40faches höher als die feldrelevante Konzentration waren, wurde festgestellt, dass die Brutzellenanzahl im Verhältnis zur Bienenanzahl verringert war. Bienen aus chronisch mit Thiacloprid eingefütterten Völkern starteten mit höherem Alter zu ihrem ersten Flug aus dem Bienenstock. Die Zeit, die die Bienen als Sammlerinnen verbrachten, änderte sich nicht. Durch Beobachtungen der Brutflächen konnte festgestellt werden, dass sich die Brut in Thiacloprid-gefütterten Völkern entsprechend der Brut in Kontrollvölkern entwickelte. Aufgrund weiterer Ergebnisse wurde eine Störung der olfaktorischen Wahrnehmung von Bienen aus Thiacloprid-gefütterten Völkern vermutet. Die akut verabreichte subletale Dosis an Thiacloprid führte zu einem erheblichen Verlust an heimkehrenden Bienen und deutet auf eine Beeinträchtigung des Orientierungs- bzw. Navigationsvermögens der Bienen hin.
In den durchgeführten Versuchen wurden sowohl direkte Auswirkungen von chronischer und akuter Aufnahme subletaler Mengen an Thiacloprid, als auch indirekte Auswirkungen auf Honigbienen beobachtet. Da teilweise erst bei hohen, nicht feldrelevanten Konzentrationen in den Versuchen Effekte beobachtet wurden, kann nur bedingt durch die Verhaltensänderung von Einzelbienen auf daraus resultierende Auswirkungen auf ein gesamtes Bienenvolk unter realistischen Bedingungen geschlossen werden.
In der Dissertation mit dem Titel „Verrechtlichung von Geschichte. Parlamentarische Debatten um die gesetzlichen Bestimmungen gegen Holocaustleugnung in Österreich und Deutschland“ wurden die strafrechtlichen Bestimmungen gegen Holocaust-Leugnung in Deutschland und Österreich untersucht. Im Vordergrund stand die Frage, wie ein historisches Ereignis mit Hilfe politischer und juristischer Terminologie so gefasst und normiert werden konnte, dass die Leugnung desselben seitdem mit Hilfe des Rechts bestraft werden kann. Dazu wurden vor allem jene parlamentarischen Vorgänge und Debatten untersucht, die der Verabschiedung der Gesetze vorausgegangen sind. Die Auswertung dieser Quellen hilft auch zu verstehen, weshalb und in welcher Form die Logik dieser Gesetze in den letzten zwanzig Jahren von anderen Staaten übernommen und auf andere historische Ereignisse ausgeweitet worden ist. Neben diesem umfangreichen empirischen Teil, der auf die jeweiligen historischen Spezifika eingeht, beinhaltet die Dissertation einen stärker analytisch ausgerichteten resümierenden Schlussteil, in dem versucht wurde, mit thesenhaften Beobachtungen das Phänomen und die ideengeschichtliche Genese der Holocaust-Leugnungsgesetze nachzuzeichnen. Diese Beobachtungen umfassen unter anderem die Bereiche Geschichtspolitik, Rechtspolitik, Sprachpolitik oder auch Wissenschaftspolitik und gehen aus unterschiedlichen Blickwinkeln der Frage nach, auf welche Weise die Gesetze begründet, legitimiert und kritisiert worden sind und immer noch werden.
Im Rahmen des FAIR Projekts sollen in den Ringbeschleunigern SIS18 und SIS100 Ionenstrahlen mit höchster Intensität beschleunigt werden. Um die Raumladungsgrenze zu erhöhen, werden dazu Ionen mit mittleren Ladungszuständen verwendet. Diese haben aber größere Wirkungsquerschnitte für Umladung in Wechselwirkungen mit im Strahlvakuum vorhandenen Restgasteilchen als hochgeladene Ionen. Kommt es zu Strahlverlusten, lösen die verlorenen Ionen am Auftreffort weitere Restgasteilchen von den Wänden des Strahlrohrs und erhöhen so lokal die Restgasdichte. Die Qualität des Vakuums ist deshalb für einen stabilen Strahlbetrieb essentiell.
Im SIS100 kommen kryogene Vakuumkammern zum Einsatz, deren Wände als Kryosorptionspumpen für Wasserstoff und Helium dienen und alle schwereren Restgaskomponenten durch Kryokondensation binden können. Um die Vorhersagegenauigkeit des abteilungsinternen Programms „StrahlSim“ zur Simulation des dynamischen Vakuums zu verbessern, wurden im Rahmen dieser Arbeit das Saugvermögen und die Pumpkapazität für Wasserstoff auf einer Edelstahloberfläche untersucht.
Dazu wurde ein UHV Teststand entwickelt und aufgebaut. Dieser besteht aus einem warmen Diagnoseteil, mit dem der Gasfluss in und aus dem kalten Teil überwacht werden kann. Im kalten Teil befindet sich eine kleine Kammer mit Edelstahlwänden, für die verschiedene Temperaturen zwischen 7 und 31 K eingestellt werden können. Diese Kammer repräsentiert ein kleines Stück kryogenes Strahlrohr. Bei verschiedenen Temperaturen und Oberflächenbelegungen wurden dort jeweils das Saugvermögen und der sich einstellende Gleichgewichtsdruck im Bereich von 4E-11 bis 2E-7 mbar gemessen. Die Gleichgewichtsdrücke bei einer bestimmten Temperatur bei wachsender Oberflächenbelegung werden als Adsorptionsisotherme bezeichnet. Sie ergeben sich aus dem Gleichgewicht von thermisch desorbierenden Teilchen und deren Readsorption. Die kalte Kammer wird umgeben von einem Kryostaten, bestehend aus thermischem Schild und Außentank. Für diesen wurde die thermische Auslegung durchgeführt, die Konstruktion erfolgte extern.
Aus dem gemessenen Saugvermögen konnte die Haftwahrscheinlichkeit berechnet werden. Sie stellte sich als im Rahmen der Messgenauigkeit als unabhängig von Belegung und Temperatur heraus. Ihr Wert liegt nahe 1 mit einer Unsicherheit bis 0,1. Da sämtliche Oberflächen in den kryogenen Bereichen als Pumpen wirken, ist dieser Wert mehr als ausreichend um die für den stabilen Strahlbetrieb nötigen Vakuumbedingungen zu erreichen und stabil zu halten.
Die Isothermen hingegen sind stark von der Temperatur abhängig. Über 18 K liegen die Gleichgewichtsdrücke bereits bei minimalen Oberflächenbelegungen in für den Strahlbetrieb nicht tolerierbaren Bereichen. Mit sinkender Temperatur können die Oberflächen immer mehr Gas aufnehmen. Doch auch bei den tiefsten vermessenen Temperaturen zwischen 7 und 8 K ist ein stabiler Strahlbetrieb nur bei Belegungen von deutlich unter einer halben Monolage, etwa 5E14 Wasserstoffmoleküle pro cm², möglich.
Diese Ergebnisse wurden in StrahlSim implementiert. Zunächst wurde der Code für die Simulation von teilweise kryogenen Beschleunigern angepasst. Die wichtigste Änderung war die Einführung von thermischer Transpiration. Sie bewirkt, dass die Restgasteilchendichte an Kalt-Warm-Übergängen auf der kalten Seite erhöht ist. Mit dieser Änderung und den implementierten Ergebnissen aus den Messungen wurden Simulationen für das SIS100 durchgeführt. Mit den Isothermen konnten die bei verschiedenen Temperaturen und Bedeckungen zu erwartenden Durchschnittsdichten berechnet werden, die wiederum bestimmend für die Strahlverluste sind. Des Weiteren wurde ein mehrwöchiger Dauerbetrieb simuliert. Es zeigt sich zunächst eine Verschlechterung der Vakuumbedingungen auf Grund der langsamen Sättigung der Oberflächen. Diese verlangsamt sich aber immer mehr und stabilisiert sich bevor zu hohe Restgasdichten auftreten. Im schlechtesten Fall sind die kryogenen Oberflächen so weit gesättigt, dass sie genauso viele Gasteilchen thermisch desorbieren wie sie adsorbieren, sie also praktisch passiv sind. Auch dann wäre die Gleichgewichtsdichte im Beschleuniger noch tief genug, um Verluste durch Umladung hinreichend niedrig zu halten.
Als problematisch könnten sich hingegen dynamische Temperaturerhöhungen der Kammerwände erweisen. In diesem Fall stellt sich praktisch verzögerungsfrei der durch die neue Isotherme definierte Gleichgewichtsdruck ein, der auch bei wenigen Kelvin Temperaturunterschied bereits um mehrere Größenordnungen höher liegen kann. Sind Temperaturerhöhungen während des Betriebs zu erwarten, sollten die Oberflächen so frei wie möglich von Wasserstoff gehalten werden. Dazu kann man sich eben diesen Effekt zunutze machen: Durch temporäres Anwärmen der Oberflächen unmittelbar vor dem Strahlbetrieb können die Oberflächen schnell von Wasserstoff befreit werden, der dann von lokalisierten Pumpen aus dem System entfernt werden kann.
Snake bite envenoming often results in disability or death of breadwinners of poor families in the rural tropics and the subtropics of Nepal. Identification of the medically relevant snake species, circumstances of venomous snake bites, prehospital care of their bites and human responses to snakes and snake bite is, therefore, crucial to enable victims or first aider to select the appropriate first aid measures, physicians to anticipate complications and to use appropriate treatment protocols as well as the local community to implement prevention strategies. Inadequate educational gaps exist in Nepal and hinder identification of snakes involved in bites. To fill this gap, I aim to provide an evidence-based list of medically relevant snake species. Snake specimens brought by patients bitten or their attendants from the tropical and subtropical regions in southeastern, southcentral, and southwestern Nepal to snake bite treatment centres over a period from 2010 through 2014, were taxonomically identified and medical records of envenoming were evaluated.
In Nepal, the epidemiology of snake bite is poorly known. Here I describe the ecological circumstances of proven krait (Bungarus spp.) and Russell´s Viper (Daboia russelii) bites to elucidate and examine, whether environmental circumstances or human behaviour contributed to envenoming. In a cross-sectional study, data about prehospital care, environmental circumstances of 46 krait and 10 Russell´s Viper bites were evaluated. Patients were interviewed using structured interview forms. Snake bite prone communities were surveyed to test people´s knowledge on snakes and their attitude towards venomous snakes in general.
Of 349 snakes involved in bites, 199 (57%) specimens were found to be medically relevant venomous snakes that included 11 species belonging to six genera and two families. Among them, Naja naja (n = 76, 22%), Bungarus caeruleus (n = 65, 19%) and Trimeresusurs albolabris (n = 10, 3%) were the most widely distributed snakes. Daboia russelii (n = 10, 3%) was found to be restricted to the southwestern part of Nepal. For B. walli, a previously poorly known species, 13 voucher specimens represent the first country records of this species as well as the first documented cases of involvement in snake bite envenoming by this species in Nepal.
Numerous snake bites (33%) occurred at night, during the rainy season, and are mainly due to Bungarus species, particularly B. caeruleus. Bites of cobras and Russell’s Vipers are a risk at daytime. Evaluation of data regarding the place where the bite happened, indicates that the snake bite risks appear to be as high in residential areas, in and around houses, as in rural areas. In cases of kraits (n = 46), 61% of the bites occurred while the victim was sleeping indoors, those of Russell´s Vipers mainly during agricultural activities in the fields. Analysis of socio-demographic data revealed that both krait and viper bites predominantly affected farmers or their family members. However, snake bites involved also people of higher socio-economic status, which suggests that it is not a health problem of poor people only living in the rural areas of Nepal.
A small number of snake bite victims (n = 7) sought help from traditional healers, but most patients went to hospitals for medical treatment using motorbikes (65%) or were transferred by ambulance cars (22%). As a first aid measure, most patients (78%) had used a tourniquet, which is of doubtful value and has often severe sequelae, instead of applying the WHO recommended pressure immobilisation bandage or local compression pad. The overall case fatality rate was calculated to be 10%, but up to 17% in cases of Bungarus spp. bites.
Rural community people were found to be extremely afraid of snakes, a major reason for indiscriminate killing of even harmless snakes, e.g., Lycodon aulicus, which were wrongly considered to be venomous. This is mainly due to the poor knowledge on snakes in general and on their role in providing ecological services, which may eventually lead to a decline in snake populations and even the extinction of rare species.
The results of the present study strongly emphasize that snake bite is an important public health issue in Nepal. There is an urgent need to improve the knowledge of people on snakes and to try changing their attitudes towards these reptiles, in addition to documenting the biodiversity and distribution of medically relevant snakes, the epidemiology and circumstances of their bites. Avoiding high-risk behaviour (e.g., killing of snakes), using screened doors and windows are some of the suggested measures preventing snake bite. Early and accurate identification of the snakes involved should help physicians to apply timely treatment, eventually referring the patient to the appropriate hospital. This also has important implications in developing public health and conservation strategies, to the benefit of the people of Nepal.
Der Pilz Podospora anserina ist seit mehr als fünf Jahrzehnten ein wichtiger Modellorganismus für die Alternsforschung. Insbesondere die Mitochondrien, essentielle eukaryotische Zellorganellen – wegen ihrer Funktion im Energiestoffwechsel häufig auch als „zelluläre Kraftwerke“ bezeichnet, sind Schlüsselfaktoren für den Alterungsprozess dieses Organismus.
Im Rahmen einer vorangegangenen Diplomarbeit wurde daher der Einfluss der mitochondrialen CLPXP-Protease, einem bisher noch wenig erforschten Bestandteil der Proteinqualitätskontrolle in Mitochondrien, auf die Alterung von P. anserina untersucht. Mitochondriale CLPXP-Proteasen sind, wie auch ihre bakteriellen Pendants, aus zwei verschiedenen Untereinheiten aufgebaut: der Protease-Komponente CLPP und der Chaperon-Komponente CLPX. Die Deletion des Gens PaClpP, kodierend für CLPP in P. anserina, führte zu einer überraschenden Verlängerung der gesunden Lebensspanne der Mutante. Darüber hinaus war es möglich, den pilzlichen PaClpP-Deletionsstamm durch Einbringen von CLPP des Menschen zu komplementieren. Dies beweist, dass die Proteasen CLPP des Menschen und von P. anserina funktionell homolog sind. Dadurch eröffnete sich die Perspektive, diesen einfachen Modellorganismus für die Gewinnung potenziell auf den Menschen übertragbarer Erkenntnisse einzusetzen. Bedeutenderweise ist die menschliche CLPXP-Protease wahrscheinlich involviert in die Entstehung verschiedener Krankheiten, darunter das Perrault-Syndrom sowie einige Krebsarten. Die zugrundeliegenden Mechanismen sind jedoch noch weitestgehend unverstanden.
Ziel des in dieser Dissertation beschriebenen Forschungsprojektes war daher die Gewinnung genauerer Einsichten in die molekulare Funktion und die daraus folgende biologische Rolle der mitochondrialen CLPXP-Protease von P. anserina. Der wohl wichtigste Punkt für das detaillierte Verständnis einer Protease ist die Kenntnis ihres Substratspektrums, d. h. der von ihr abgebauten Proteine. Tatsächlich wurde aber bis heute noch in keinem eukaryotischen Organismus eine umfassende Analyse der Substrate einer mitochondrialen CLPXP-Protease vorgenommen. Um diese Wissenslücke zu füllen, wurde in der vorliegenden Arbeit eine ursprünglich in Bakterien entwickelte Verfahrensweise, der sogenannte CLPP „Substrat-trapping Assay“, in P. anserina implementiert. Dafür mussten zunächst die notwendigen handwerklichen Voraussetzungen für den Assay geschaffen werden, insbesondere die effiziente Affinitätsaufreinigung von Proteinen aus isolierten Mitochondrien – einer bisher in P. anserina noch nicht angewandten Technik. Unter Verwendung verschiedener neu hergestellter Varianten der menschlichen Protease-Komponente CLPP, darunter einer proteolytisch inaktiven Variante zum „Einfangen“ von Substraten, konnte der CLPP „Substrat-trapping Assay“ in P. anserina erfolgreich durchgeführt werden. Insgesamt wurden, in Kooperation mit der Arbeitsgruppe von Julian D. Langer (Max-Planck-Institut für Biophysik; Durchführung von massenspektrometrischen Analysen) nahezu 70 spezifische Proteine erstmalig als potenzielle Substrate oder Interaktionspartner einer mitochondrialen CLPXP-Protease identifiziert. Bei einem Großteil dieser Proteine handelt es sich um Enzyme und Komponenten verschiedener Stoffwechselwege – vor allem um solche, die eine zentrale Rolle im mitochondrialen Energiestoffwechsel spielen. Die Ergebnisse der vorliegenden Arbeit legen somit folgende Arbeitsthese als Schlussfazit und gleichzeitig Ausganspunkt für zukünftige Untersuchungen nahe:
Die hauptsächliche molekulare Funktion der mitochondrialen CLPXP-Protease in P. anserina ist die Degradation von Stoffwechselenzymen und ihre biologische Rolle demnach die Kontrolle und Aufrechterhaltung des mitochondrialen und zellulären Energiestoffwechsels.
Insgesamt ist die auf Grundlage des CLPP „Substrat-trapping Assay“ in P. anserina anzunehmende Rolle der mitochondrialen CLPXP-Protease als regulatorische Komponente des mitochondrialen Energiestoffwechsels erstaunlich gut mit Beobachtungen in anderen eukaryotischen Organismen, gerade bezüglich der Relevanz der CLPXP-Protease des Menschen für diverse Krankheiten, zu vereinbaren. Somit erscheint es überaus sinnvoll und vielversprechend, dass in dieser Doktorarbeit erstellte und bisher beispiellose Kompendium potenzieller in vivo Substrate und Interaktionspartner dieser Protease auch als Referenz für zukünftige Untersuchungen außerhalb von P. anserina anzuwenden.
In dieser Arbeit werden Projekte beschrieben, in denen das Adsorptionsverhalten von Proteinen und Bakterien an verschiedene Materialoberflächen manipuliert wird.
Durch die Reaktion verschiedener oxidischer Oberflächen mit Glycidol konnten biorepulsive Polyglycerolschichten erzeugt werden. Für die Herstellung dieser Polyglycerolschichten wurden zwei unterschiedliche Verfahren entwickelt und untersucht. Die erste Methode beruht auf der Bildung einer aminoterminierten Monolage auf Silicium-Oberflächen, an der in einem zweiten Schritt die Polymerisation von Glycidol durchgeführt wird. Die Dicke der angebundenen Polyglycerolschicht ist abhängig von der Beschichtungsdauer, wobei die dicksten Schichten bis zu 98% der Bakterienadhäsion unterdrücken können. Das zweite Verfahren ist die direkte Anbindung von stabilen Polyglycerol-Beschichtungen an Silicium-, Aluminium- oder Stahl-Oberflächen. Je größer die abgeschiedene Polyglycerolmenge ist, desto höher ist die Biorepulsivität der Schicht, was durch Adsorptionstests mit Proteinen und ermittelt wurde.
Polyglycerolschichten eignen sich besonders gut für die nachträgliche Modifizierung. So konnten beispielsweise mittels Elektronenstrahlen laterale Strukturierungen der Polyglycerol-beschichteten Oberflächen erfolgreich durchgeführt werden. Sensorisch aktive Moleküle wie Ethylendiamintetraessigsäure oder Biotin konnten im Rahmen dieser Arbeit nachträglich an Polyglycerolschichten angebunden werden. Die Aktivität der Bindungsstellen nach der Anbindung an die Oberfläche konnte dabei durch spezifische Erkennungsereignisse nachgewiesen werden.
Im zweiten Teil dieser Arbeit wurden selbstanordnende Monoschichten mit Oligoethylenglycol (OEG)-Kopfgruppen und Thiolat-Ankergruppen verwendet, um lateral strukturierbare, biorepulsive Schichten auf Gold zu erzeugen. Es wurde untersucht, ob derartige OEG-Monolagen kontrolliert durch langwelliges UV-Licht (390 nm) abgebaut werden können, um proteinbindende und proteinrepulsive Bereiche auf einer Substrat-Oberfläche zu generieren. Die Bestrahlung mit UV-Licht bewirkte die Oxidation und Abspaltung der Ethylenglycol-Einheiten, wodurch die unspezifische Adsorption von Proteinen erfolgen kann. Zusätzlich konnten Photooxidations-Reaktionen an der Thiolat-Ankergruppe nachgewiesen werden, welche die Ablösung des SAM-Bausteins zur Folge haben.
Für den Einsatz von Lithographie-Techniken in mikrofluidischen Anlagen wurde das Abbauverhalten der biorepulsiven Monolage bei der Bestrahlung unter Wasser untersucht. In Abwesenheit von molekularem Sauerstoff kommt es hier lediglich zur Spaltung der Etherbindung zwischen den Ethylenglycol-Einheiten. Die Beobachtung, dass die An- bzw. Abwesenheit von molekularem Sauerstoff zu zwei unterschiedlichen Abbaumechanismen führt, kann für die Feinabstimmung der Oberflächenbeschaffenheit und somit der Proteinanlagerung genutzt werden.
Biorepulsive OEG-Monolagen können auch dazu verwendet werden, um gezielt bestimmte Biomoleküle anzulagern. Dazu können die Monolagen mit Erkennungsstellen ausgestattet werden, welche die spezifische Anbindung einer Biomolekül-Spezies ermöglichen. Gerade bei der Detektion von großen Biomolekülen oder Mikroorganismen spielt jedoch nicht nur die chemische Zusammensetzung, sondern auch die Ausrichtung der Bindungsstelle eine entscheidende Rolle. Für die Untersuchung des Orientierungseinflusses wurden Moleküle verwendet, die neben einer Mannose-Einheit als Bindungsstelle für Bakterien auch eine Azobenzol-Gruppe, welche die strahlungsinduzierte reversible Schaltung der Konformation ermöglicht, tragen. Bakterien-Adhäsionstests zeigten, dass sich die Orientierung der Mannose-Einheit auf die Anbindung der Bakterien auswirkt.
Im Rahmen dieser Arbeit wurden neuartige Methoden zur Herstellung, Charakterisierung und Strukturierung biorepulsiver und biosensorischer Schichten entwickelt. Die dadurch gewonnenen Erkenntnisse sind von bedeutender wissenschaftlicher Relevanz und ermöglichen die potentielle industrielle Anwendung der entwickelten Methoden im Kontext der Material- und Biotechnologie sowie der Nanofabrikation.
Barriereinseln und Nehrungshaken, geformt durch eine Kombination aus Wind, Wellen, Strömung und Küstenlängstransport gelten als geologisch junge und morphologisch hoch aktive Küstenbereiche und variieren häufig in Ursprung, Genese und Entwicklung. Bisherige Untersuchungen zur Stratigraphie dieser durch engräumige Fazieswechsel geprägten Sedimentationsräume basieren häufig allein auf Bohrungen. Daher sind die interne sedimentologische Struktur und die Prozesse, die zur Entwicklung von Barriereinseln und Nehrungshaken geführt haben, oftmals unzureichend untersucht.
Ziel der folgenden Studie war es, anhand hochauflösender Georadarmessungen (GPR) und Bohrungen die Entstehung und interne sedimentäre Architektur der Nehrungshaken von Sylt und Amrum zu rekonstruieren. Durch die Korrelation von Sediment- und Radarfazies konnten über den bisherigen Kenntnisstand hinaus wertvolle, sich ergänzende Informationen zur Geologie des oberflächennahen Untergrundes der Inseln Sylt und Amrum gewonnen werden. Auf dieser Grundlage wurden für den Süden Sylts, die Westküste von Amrum und den Norden Amrums stratigraphische Modelle entwickelt, die schematisch den sedimentologischen Aufbau, die geomorphologisch-geologische Genese und die am Aufbau beteiligten Prozesse zusammenfassen. Die Einordnung in einen absoluten Zeitrahmen wurde durch Datierungen (AMS Radiokohlenstoff-Methode, Aminosäure-Racemisierungs-Methode) ermöglicht.
Die Ergebnisse sind als Beispiel für die Heterogenität und Individualität von Nehrungshaken und Barriereinseln zu sehen. Es konnte gezeigt werden, dass Nehrungshaken hinsichtlich ihres Aufbaus, ihrer sedimentären Struktur sowie den Prozessen ihrer Entstehung sehr unterschiedlich sind.
Mit Ausnahme der heutigen Dünen zeigt die Stratigraphie der Ansatzzone des südlichen Nehrungshakens von Sylt an den zentralen Inselgeestkern südlich der Ortschaft Rantum insgesamt eine Vergröberungs-Sequenz von tonigem Mischwatt im Liegenden über Sandwatt bis zu den gröberen washover-Schichten im Hangenden. Eine Besonderheit und regionale Abweichung stellt die Stratigraphie bei Puan Klent/Thörnhörn dar. Dort wurde im Liegenden der Watt-Fazies ein fossiler Strandhaken nachgewiesen.
Die transgressive Sequenz der Ansatzzone des südlichen Nehrungshakens von Sylt bei Rantum auf Sylt ist kennzeichnend für eine landwärts gerichtete Wanderungsbewegung der Inselbarriere unter steigendem Meeresspiegel. Das Resultat des als „barrier rollover“ bezeichneten morphologischen Prozesses ist die allmähliche Erosion der westlichen Barrierefront. Das erodierte Sediment der Westküste wurde entweder durch overwash-Prozesse im rückwärtigen Inselbereich als washover fan abgelagert oder gelangte in den Küstenlängstransport, wurde zur Inselspitze transportiert und dort akkumuliert. Diese führte allmählich zum Wachstum und zur Progradation der südlichen Inselspitze (Hörnum Odde) in die sich südlich anschließende Tiderinne des Hörnum-Tiefs. Zahlreiche Erosionsdiskordanzen unterhalb des heutigen Meeresspiegels markieren ursprüngliche Inselspitzenendpositionen (time lines) und sind auf hochenergetische Sturmflutereignisse zurückzuführen. Die time lines werden nach Süden hin jünger und zeichnen unterschiedliche Stadien der Progradation nach, die ein episodisches Wachstum der Inselspitze in Abhängigkeit von der Sedimentverfügbarkeit und der Sturmflutintensität belegen. Alte Erosionsdiskordanzen belegen die frühe submarine Genese der Hörnum Odde. Ein starkes Wachstum der südlichen Inselspitze Sylts lässt sich im Mittelalter nachweisen.
Sturmfluten führten zudem immer wieder zu overwash-Prozessen und Überschwemmungen der noch jungen Inselspitze. Dies belegen ältere washover-Ablagerungen, die in unterschiedlichen Phasen der Nehrungshakenentwicklung nachgewiesen werden konnten. Rezente overwash-Prozesse sind auf die heutigen Dünen beschränkt und wurden in dieser Arbeit mit der Fragestellung untersucht, ob und inwieweit das Georadar als Methode zur Prospektion von washover-Sedimenten herangezogen werden kann.
Die unmittelbare Nachbarschaft der beiden Nehrungshaken von Sylt und Amrum lässt einen ähnlichen sedimentären Aufbau, eine analoge Genese in einem einheitlichen Prozess-System sowie ein vergleichbares Alter vermuten. Durch die Untersuchungen im Norden von Amrum sollte festgestellt werden, in wie weit das für die Hörnum Odde nachgewiesene Zusammenspiel von Küstenlängstransport, Progradation, sturmflutbedingten overwash-Prozessen und die zeitliche Unterbrechung des Längenwachstums durch Sturmfluterosion auch für Amrum zutrifft. Entgegen der Erwartungen unterscheidet sich der nördliche Nehrungshaken der Insel Amrum signifikant vom südlichen Nehrungshaken der Nachbarinsel Sylt.
Studien zur Landschaftsentwicklung an der Westküste Amrums wurden in einem Modell zusammengefasst, dass die Sedimentationsbedingungen, die im Westküstenvorfeld herrschten bevor der Kniepsand an die Insel heranwanderte, beschreibt. Auf der Landoberfläche des ertrinkenden saaleeiszeitlichen Geestkerns wurden zu Beginn der Flandrischen Transgression feinkörnige Sedimente eines Misch- und Schlickwatts abgelagert. Es ist davon auszugehen, dass der damals noch weit vor der Küste Amrums liegende Kniepsand eine Barriere bildete und so an der heute hochenergetischen Westküste für strömungsberuhigte Sedimentationsbedingungen sorgte. Ein weiteres Modell beschreibt den Andockmechanismus des Kniepsandes an die Insel Amrum. Durch die Anlagerung des ehemaligen Außensandes und den damit einhergehenden Sedimentinput wurden die Bedingungen für eine großflächige Dünenbildung geschaffen.
Im Rahmen der vorliegenden Arbeit wurde einerseits der Einsatz lichtaktivierbarer Oligonukleotide zur Kontrolle der Leitfähigkeit entlang von DNA untersucht sowie neue photoaktivierbare Verbindungen für die Peptidchemie und für eine neu entwickelte Variante des SELEX (Systematic Evolution of Ligands by EXponetial enrichment) Verfahrens synthetisiert.
DNA vermittelte Ladungsübertragung verläuft entlang des gestapelten π-Systems der heteroaromatischen Nukleobasen. Die Leitfähigkeit von Oligonukleotiden reagiert daher empfindlich auf Störungen in der Watson-Crick-Basenpaarung. Die in der Arbeitsgruppe Heckel etablierte Technik, Nukleobasen an für die Basenpaarung relevanten Positionen mit photolabilen Schutzgruppen zu modifizieren, sollte daher mit Systemen der Ladungsübertragung in DNA kombiniert werden. Im Verlauf dieses Projekts wurden zwei literaturbekannte Varianten, in denen Ladungstransport über einen lichtinduzierten Redoxprozess zwischen Metallkomplexen ablaufen und über eine dabei unterdrückte Fluoreszenz optisch verfolgt werden sollte, als ungeeignete Systeme identifiziert. Durch den Wechsel zu elektrodengestützter Leitfähigkeitsmessung konnte der prinzipielle Effekt von Leitfähigkeit in perfekt gepaarter DNA und deutlich reduziertem Stromfluss in Oligonukleotiden mit Fehlpaarungen gezeigt werden. Beim Einsatz photolabil geschützter Oligonukleotide konnte jedoch auch in diesem System noch nicht der gewünschte Effekt gefunden werden.
Im zweiten Projekt dieser Arbeit wurden neue photolabile Verbindungen hergestellt, die Peptide nach ihrem Einbau in das Peptidrückgrat durch Zwei-Photonen-Anregung mit IR-Licht spalten sollen. Drei entsprechende Nitrodibenzofuran-Verbindungen und ein Cumarin-Baustein konnten erfolgreich synthetisiert werden. Die neuen Moleküle zeigten im Rahmen der Peptid-Festphasensynthese Stabilitätsprobleme. Diese Schwierigkeiten konnten durch Peptid-Kopplungen in Lösung umgangen werden. Mit Hilfe eines der hergestellten Bausteine wurden zwei Tripeptide hergestellt, die jeweils mit dem Farbstoff ATTO565 markiert und hinsichtlich ihrer photochemischen Eigenschaften charakterisiert wurden. Der neue Baustein zeigte neben den Eigenschaften als photospaltbare Gruppe, dass er gleichzeitig ein Quencher für den Farbstoff ATTO565 darstellt. Nach Belichtung stieg die Fluoreszenz um den Faktor 81 an. Die Aktivierung gelang wie erwartet mit Ein- und Zwei-Photonen-Anregung. In Kollaboration mit der Arbeitsgruppe von Prof. Heilemann konnten Antiköper mit einem der Tripeptide modifiziert werden und die Kompatibilität der Verbindung mit hochaufgelöster Einzelmolekül-Fluoreszenzmikroskopie demonstriert werden.
Im letzten in dieser Arbeit thematisierten Projekt wurden neue lichtspaltbare Verbindungen für eine Variante des SELEX-Prozesses hergestellt. Diese Verbindungen erlauben die temporäre Einführung einer Indol Modifikation an Alkin-modifizierte Oligonukleotide über die sogenannte Click-Chemie. Neue chemische Modifikationen wie die hier verwendeten Indole erhöhen die chemische Vielfalt der Oligonukleotide. Eine größere Vielfalt führt zu neuen potentiellen Wechselwirkungen gegenüber Verbindungen, gegen die mit Hilfe herkömmlicher SELEX-Verfahren keine Aptamere erzeugt werden konnten. Da die chemische Modifikation über eine photolabile Gruppe an die Oligonukleotide gebunden wird, kann sie photochemisch von der DNA gespalten werden, wodurch eine Interferenz der Modifikation mit den enzymatisch katalysierten Schritten innerhalb der SELEX ausgeschlossen werden kann.

Although there are continuous attempts to differentiate between primary and secondary metabolites, the borders often blur in the eye of almost perfect interconvertability of all such matter. With chemistry and physics dominating this domain of biology it is an interdisciplinary endeavor to tackle the questions surrounding the workings of the metabolic pathways involved, searching for answers that ultimately help us to better understand life and find solutions to problems that affect us humans. One area of biochemistry that serves as a formidable example of the intertwined primary and secondary metabolic pathways are fatty acids, essential components of bacterial membranes, sources of energy and carbon but also important building blocks of several natural products. The second area to be mentioned is the metabolism of amino acids, the basic components of proteins and enzymes, which also serve as precursors to a diverse set of metabolites with many biological purposes.
This work focuses on these two areas of biochemistry, as several intermediates of their metabolism serve as building blocks for complex secondary metabolites whence many interesting and bioactive natural products are derived. The powerful and relatively novel tool of click-chemistry is employed to track azide-labeled precursors of primary and secondary metabolism in various bacterial strains to observe biochemistry at work and adds to the knowledge gained through other methods. The methods presented in this work serve the observation of fatty acid biosynthesis, degradation, modification and transport through direct ligation of azido fatty acids with cyclooctynes on one hand, leading to a revision of fatty acid transport in general. On the other hand a cleavable azide-reactive resin is devised to generally track the fate of azidated compounds through the myriads of metabolic pathways offered by entomopathogenic bacteria possessing a rich secondary metabolism. The resulting findings led to the identification of several antimicrobial peptides, amides and other compounds of which many had remained so far undetected in the strains that underwent investigation, underlining the worth of this method for future metabolomic research and beyond.

The mammalian family of bears (Ursidae) comprises eight extant species, occurring on four different continents. Among them are the iconic and well-known brown and polar bears, both widely distributed across the Northern hemisphere. Their intraspecific genetic structuring has been extensively investigated, albeit with a focus on genetic markers from maternally inherited parts of their genomes (mitochondrial DNA). The evolutionary relationship and divergence time between brown and polar bears have recently triggered an extensive debate, while less focus has been put on to other parts of the ursid phylogeny, particularly to a clade of three Asian bear species. To date, whole genomes of more than 100 bear individuals from four different species have been sequenced. Yet, one fundamental part of the genome has been largely omitted from specific analyses, in bears as well as in most other mammals: the Y chromosome.
The mammalian Y chromosome provides a unique perspective on the evolutionary history of organisms due to its distinct features, and specifically reflects the patriline because of its male-specific inheritance. The characteristics of this chromosome make it well suited to complement and contrast evolutionary inferences based on other genetic markers, and to uncover processes like sex-biased gene flow and hybridization. The unique insights that can be gained from analyses of Y-linked genetic variation made me utilize this part of the genome to investigate the evolution of male lineages in bears. Studying the patriline is particularly promising in this taxonomic group because of male-biased dispersal and a complex and fast radiation of bears. The analysis of Y-chromosomal genetic markers is thus the common theme of this dissertation: I present the identification of large amounts of Y-chromosomal sequence, the development of male-specific markers from such sequences, and the application of these markers to trace the evolution of male lineages of different bear species.
Specifically, I developed a molecular sex determination system based on the detection of two Y-linked fragments that allows to reliably discriminate between females and males from seven different bear species (Bidon et al. 2013). The approach is highly sensitive, bear-specific, and can be applied in standard molecular laboratories. This makes it valuable in conservation genetics and forensic applications, e.g. to analyze non-invasively collected samples.
Furthermore, I used Y-linked markers in a comprehensive and range-wide sample of brown and polar bears, and show that male-biased gene flow plays an important role in distributing genetic material throughout the ranges of both species (Bidon et al. 2014). In brown bears, I detected a lack of paternal population structuring which is in strong contrast to the detailed structuring of the matriline.
Analyzing Y-chromosomal sequences from all eight bear species, I present a phylogeny of the patriline that largely resembles the topology from other nuclear markers but is different from the topology of the mitochondrial gene tree (Kutschera et al. 2014). This discordance among loci generates interesting hypotheses about inter-species gene flow, particularly among American and Asiatic black bears.
With the identification of almost two million basepairs of Y-chromosomal sequence and the analysis of an unprecedented large male-specific dataset in polar bears, a high-resolution view on the distribution of their intraspecific variation was obtained (Bidon et al. 2015). In particular, two clades that are divergent but do not show pronounced phylogeographic structure were detected, confirming the great dispersal capacity of males of this high arctic species.
This dissertation thus represents a comprehensive investigation of Y-linked genetic variation on the intra- and interspecific level in a non-model organism. With my research, I contribute to an increased understanding of the complex evolutionary history of bears. In particular, I show that male-biased gene flow strongly influences the distribution of nuclear genetic variation, and that the contrast between phylogenies of differentially inherited markers can help to understand interspecific hybridization between closely related species. Moreover, my findings demonstrate the potential of Y-chromosomal markers to uncover unknown evolutionary patterns and processes. This applies not only to bears but to many species, even such that are generally well known and well described.
In this work, the complex structure of ionization and dissociation pathways on the potential energy curves in small molecules were investigated that are initiated by the absorption of a sequence of multi-color pulses in the XUV, VUV, and IR spectrum. Femtosecond pump-probe spectroscopy was used to track the evolution of nuclear dynamics in neutral hydrogen molecules. Previously unpublished excitation and ionization pathways leading to the dissociative ionization of hydrogen molecules were investigated by employing 3D momentum imaging spectroscopy. These studies were extended to oxygen molecules where an XUV attosecond pulse train coherently ionized several electronic states of O2+ followed by the dissociation of the molecule via multiple pathways. The infrared electric field of the driving laser was then used to couple the electronic and nuclear wave-packets, thus, manipulating the dissociation dynamics of the molecule on an attosecond time scale.
In order to perform the experiments presented here, a novel experimental setup was developed and constructed. It combines an existing high-flux High Harmonic Generation light source that delivers attosecond pulse trains in the VUV and XUV spectrum with a state-of-the-art 3D momentum imaging apparatus (COLTRIMS), as well as a beamline consisting of several experimental tools enabling the selection, characterization, and propagation of the photon spectrum.
Cyanobacteria belong to the most widely distributed microorganisms in the biosphere and contribute significantly to global primary production. Their metabolism is based on oxygenic photosynthesis and some cyanobacteria can fix elemental nitrogen. Obligate photosynthetic diazotrophs have a particularly high iron demand in comparison to heterotrophic bacteria. Nevertheless the understanding of iron acquisition in cyanobacteria is just beginning to emerge. Iron acquisition in bacteria comprises highly specific transport of siderophore-iron complexes over the outer membrane by TonB-dependent transporter (TBDT). The transport itself is active and energized by a multi-complex localized to the inner membrane termed the TonB-system (TonB-ExbB-ExbD). The siderophore-iron complexes are further transported into the cytosol by a binding protein dependent ABC-transporter. Cyanobacterial iron acquisition response has most extensively been studied in unicellular, non-siderophore synthesizing cyanobacteria in the genus Synechococcus and Synechocystis. Anabaena sp. PCC 7120, however, is a different model organism as it is a freshwater living, siderophore synthesizing and, truly multicellular microorganism. It can be assumed that siderophore synthesis and siderophore-dependent iron uptake are tightly coordinated processes, therefore Anabaena represents a different model organism as compared to non-siderophore producing cyanobacteria. Moreover the surprisingly abundant protein family of 22 putative TBDTs in Anabaena indicates a high complexity of TonB-dependent uptake systems. Sequence similarity analysis revealed 4 putative tonB encoding genes (alr0248, all3585, all5036, alr5329), 2 putative exbB-exbD encoding gene cluster (alr0643-alr0644, all5047-all5046), one single standing putative exbB encoding gene (alr4587) and several hypothetical binding-protein-dependent ATP binding cassette (ABC)-type transporter encoding genes (fhu-, fec- and fut-type transporter).
In this study the respond of the predeicted systems to iron-limiting conditions was analysed by qRT-PCR. The expression analysis revealed on the one hand an enhanced transcription of all5036 (tonB3), all5047-all5046 (exbB3-exbD3) and the fhu-like encoding genes (all0387-all0389) under iron-limitation and at the same time down-regulation of expression under enhanced iron concentrations. Summerizing the transcription profile of the tonB3- and the fhu-system showed an expression regulated by iron-availability. To further characterize the role of TonB3-, ExbB3- and the Fhu-system, mutants thereof were generated. None of the generated mutants, except for the exbB3 mutant, could be fully segregated, suggesting an essential character of the genes. Characterization of the mutants revealed enhanced expression of iron-starvatrion indicator genes (isiA, fhuA) and altered growth of the tonB3 mutant under iron-limiting conditions. The iron starvation phenotype was further strengthened by enhanced siderophore secretion in the tonB3, exbB3 and fhuC mutants. Taken as a whole the results strongly indicate involvement of the tonB3- and the fhu-system in siderophore-dependnet iron uptake in Anabaena.
Investigation of the tonB2 (all3585) mutant under iron and citric acid limitation resultated in altered growth of the mutant. However, growth could be restored by addition of iron chlorid. Therefore a connection of the TonB2 protein to iron uptake is implied and further supported by ressitance to toxic iron concentrations. Lastly, mutation of tonB1 (alr0248) reuslted in insensibility to toxic manganese and copper concentrations and macrolid antibiotics. The altered permeability of the outer membrane may be a result of decreased expression of seven putative porin encoding genes in the mutant. A possible role in transcriptional regulation of porin expression is discussed.
This dissertation examines the language of politics of leading figures of the ex-Free Aceh Movement or Gerakan Aceh Merdeka‟ (GAM) leading figures in selected Aceh media during the 2012 gubernatorial election campaign. By analyzing their symbolic acts, topic selection, campaign methods, and campaign locations as reported in five selected Aceh media, I demonstrate the process of ex-GAM political rhetoric in the post-conflict election setting. The mixed method approach used in the dissertation includes the following: grounded, content analysis, case study, and rhetorical analysis. Data were collected from three local daily broadsheet newspapers (Serambi Indonesia, Rakyat Aceh and Harian Aceh) and two online news portals (The Atjeh Post and The Globe Journal). The research found that ex-GAM political rhetoric was deeply divided into two opposing political camps: the Irwandi camp as the incumbent independent and the PA camp as the challenger from a newly-established local political party. Both camps highlighted the importance of expressive symbolic acts, such as explaining and making promises in their campaigns. Irwandi introduced more varied, specific, practical, and concrete topics in its rhetoric with diverse and less formal campaign methods that appealed to those who envision a prosperous future in Aceh. By contrast, PA employed general, unrealistic, and abstract topics in its rhetoric with less attractive and more formal campaign methods that appealed to emotional and historical romanticism. In relation to ex-GAM political rhetoric and campaign locations, both focused on the traditional regions in northeast Aceh or in the conflict-sensitive regions where most of their loyalists reside, and on big regencies/cities where the potential voters live. However, during the campaign Irwandi appeared less frequently in ethnically diverse locations, whereas PA appeared more in non-traditional regions such as the previously isolated and less-developed regions in the central highlands and southeast regions of Aceh. In this way they highlighted the rhetoric of economic and infrastructure development and that of unity in diversity taking into account the multi-ethnicity of the voters. In terms of overall media appearances, except in The Atjeh Post, where Muzakir Manaf from PA was dominant, Irwandi Yusuf was the most-reported ex-GAM during the election campaign in the selected media. Finally, the available evidence seems to suggest that Irwandi‘s language of politics was rhetorically more attractive than that of other ex-GAM leaders.
Cardiac progenitor cells hold great potential for regenerative therapies in heart disorders. However, the molecular mechanisms regulating cardiac progenitor cell expansion and differentiation remain poorly defined. Here we show that the multi- adaptor protein Ldb1, which mediates interactions between different classes of LIM domain transcription factors, is a multifunctional regulator of cardiac progenitor cell differentiation. Ldb1-deficient embryonic stem cells (ESCs) show a markedly decreased expression of second heart field (SHF) marker genes and subsequently impaired cardiomyocyte differentiation. Conditional ablation of Ldb1 in the early SHF using an Isl1-Cre driver led to embryonic lethality at Embryonic day (E)10.5 with cardiac abnormalities including a significantly smaller right ventricle and a shortened outflow tract, supporting a crucial role of Ldb1 in the SHF. Mechanistically we show that the importance of Ldb1 for SHF development is two-fold: On the one hand, Ldb1 binds to Isl1 and protects it from proteasomal degradation, as a consequence of which Ldb1-deficiency leads to an almost complete loss of Isl1+ cardiovascular progenitor cells. On the other hand the Isl1/Ldb1 complex promotes long-range promoter-enhancer interactions at the loci of the core cardiac transcription factors Mef2c and Hand2. Chromosome conformation capture followed by sequencing (3C- seq) identified specific Ldb1-mediated interactions of the Isl1/Ldb1 responsive Mef2c anterior heart field enhancer with genes which play key roles in cardiac progenitor cell function and cardiovascular development. These interactions are of critical importance to regulate the expression of the downstream target genes since their expression levels are strongly dependent on the Ldb1/Isl1 levels. Overexpression of an Ldb1 mutant, which contains the LIM interaction domain and thereby can protect Isl1 protein from degradation, but lacks the dimerization domain and thus cannot promote long-range interactions, does not collaborate with Isl1 to regulate the expression of their common targets and results in defects in Isl1+ cardiac progenitor differentiation. In this thesis we show one of the first examples of genome-wide chromatin reorganization mediated by a developmental regulated, cell type specific, transcription complex. Ldb1 in concert with Isl1 promotes long range promoter- enhancer and enhancer-enhancer interactions in order to create active chromatin hub where gene important for heart development can be co-regulated. Moreover, Isl1 and Ldb1 genetically interact during heart development, as Isl1/Ldb1 haplodeficient embryos show various cardiac anomalies. The dosage-sensitive interdependence between Isl1 and Ldb1 in the expression of these key factors in cardiogenesis, further supports a key role of the Isl1/Ldb1 complex in coordinating a three dimensional genome organization, upstream of a regulatory network driving cardiac differentiation and heart development.
In conclusion, the Isl1/Ldb1 complex orchestrate a genome-wide three dimensional chromatin reorganization resulting in a transcriptional program responsible for the differentiation of multipotent cardiac progenitor cells into cardiomyocytes.
Eye-Tracking bezeichnet das Messen und Aufzeichnen der Blickbewegungen einer Person. Historisch gesehen basiert Eye-Tracking auf Beobachtungen des Testleiters, der das Blickverhalten der Probanden während des Versuchsablaufes oder die Videoaufzeichnung des Blickverhaltens eines Probanden in einer Testsituation kodierte. Dabei konnte allerdings nur die Blickrichtung des Probanden erhoben werden. Heutzutage ist es jedoch möglich, aufgrund neuerer, automatisierter Eye-Tracking-Techniken detailliertere Blickbewegungen, wie z.B. Fixationen und Sakkaden, zu messen. Diese Verbesserung der Eye-Tracking-Technik ermöglicht nicht nur passives Eye-Tracking, sondern auch aktives Blickkontingenz-Eye-Tracking. Passives Eye-Tracking bezeichnet das Messen und Aufzeichnen des Blickverhaltens, um herauszufinden, wo der Proband hinschaut. Im Gegensatz dazu erhebt das aktive Blickkontingenz-Eye-Tracking nicht nur, wo ein Proband hinschaut, sondern ermöglicht dem Probanden auch, die Stimuli, die auf einem Bildschirm präsentiert werden, aktiv zu verändern oder zu kontrollieren. Dabei wird das Blickverhalten online kodiert, und spezifisches Blickverhalten ist an eine kontingente Veränderung der Stimuli auf dem Bildschirm gekoppelt. Deshalb kann das aktive Blickkontingenz-Eye-Tracking eingesetzt werden, um den Probanden aktive Kontrolle über ihre visuelle Umwelt zu ermöglichen.
In der psychologischen Forschung ist Eye-Tracking ein wichtiges Forschungs-instrument, da das Blickverhalten in spezifischen Eye-Tracking-Aufgaben genutzt werden kann, um Aufschluss über kognitive Prozesse, wie z.B. Aufmerksamkeit, Lernen und Gedächtnis, zu gewinnen. Unterschiedliche passive und aktiv-blickkontingente Eye-Tracking- Aufgaben wurden entwickelt, um eine Vielzahl an kognitiven Prozessen im Erwachsenen- und Säuglingsalter zu untersuchen. Diese Aufgaben sind besonders wichtig in der Säuglingsforschung, da es in diesem Alter schwierig ist, kognitive Prozesse zu untersuchen. Dies hängt damit zusammen, dass es sich um eine präverbale Stichprobe, die nur über ein limitiertes motorisches Repertoire verfügt, handelt. Obwohl kognitive Prozesse von Erwachsenen anhand verbaler oder anderer motorischer Aufgaben untersucht werden können, werden passive und aktiv-blickkontingente Eye-Tracking-Aufgaben regelmäßig in dieser Altersgruppe eingesetzt, da sie zusätzliche Informationen über kognitive Prozesse liefern können. Neben der Möglichkeit zur Untersuchung von kognitiven Prozessen bieten aktiv-blickkontingente Eye-Tracking-Aufgaben den Probanden auch die Gelegenheit, ihre visuelle Umwelt aktiv zu kontrollieren. Dennoch werden aktiv-blickkontingente Eye-Tracking- Aufgaben nur selten eingesetzt, um Probanden visuelle Kontrolle über ihre Umwelt zu verschaffen.
Bis jetzt wurden aktiv-blickkontingente Eye-Tracking-Aufgaben zur Kontrolle der visuellen Umwelt nur bei Erwachsenen, aber noch nicht bei Säuglingen eingesetzt. Da diese Aufgaben jedoch auch für Säuglinge und Kleinkinder geeignet sind, besteht die Möglichkeit, diese Methode über die gesamte Lebenspanne hinweg anzuwenden. Somit kann das Erlernen des Kontrollierens der Umwelt durch Blickverhalten über die gesamte Lebensspanne untersucht werden.
Die vorliegende Dissertation hat sich genau dies zum Ziel gesetzt. Um dieses Ziel zu erreichen, wurde eine neue aktiv-blickkontingente Eye-Tracking-Aufgabe entwickelt, die sogenannte gaze-contingent learning task (GCLT). Im Wesentlichen ist die GCLT eine operante Konditionierungsaufgabe, bei der sich Probanden Kontrolle über ihre visuelle Umwelt aneignen, indem sie eine bestimmte blickkontingente Assoziation zwischen ihrem Blickverhalten und einem visuellen Effekt erlernen. Die in dieser Dissertation verwendete GCLT umfasst zwei Hauptversionen: zum einen die sog. one disc GCLT, und zum anderen die two discs GCLT. In der one disc GCLT wird ein Kreis auf der rechten Bildschirmseite gezeigt. Jedes Mal, wenn der Proband auf diesen Kreis schaut, erscheint ein Stimulus auf der linken Bildschirmseite. Somit kommt dem Kreis eine Schalterfunktion zu. In der two discs GCLT ist sowohl am rechten als auch am linken Bildschirmrand ein Kreis zu sehen. Hier kommt nur jeweils einem der beiden Kreise die Schalterfunktion zu. Um ihre visuelle Umwelt zu kontrollieren, müssen Probanden innerhalb der one disc GCLT die blickkontingente Assoziation zwischen ihren Blicken auf den Kreis und dem Erscheinen eines Stimulus erlernen, während sie in der two discs GCLT außerdem noch lernen müssen, zwischen dem Kreis mit und dem Kreis ohne Schalterfunktion zu unterscheiden.
This thesis develops a conceptual framework for a better understanding of the impact of slow-onset climate and environmental changes on human migration in developing countries. Its regional focus is on the West African Sahel, where the majority of the population depends on agriculture and thus is highly vulnerable to environmental changes. Migration from fragile environments is predominantly considered one of several household strategies to adapt to and minimise the risk of environmental stress. Based on qualitative and quantitative data from two selected rural study areas, Bandiagara in Mali and Linguère in Senegal, this thesis analyses the drivers of migration from the two areas.
The findings illustrate that, even though people highly depend on the natural environment, migration motives are manifold and that migration often is not a household strategy to cope with environmental changes. Although environmental conditions shape migration in the region and the migrants’ support is crucial for most households, environmental stress plays a relatively small role as a driver of migration - at least in Mali, where it is considerably less important than in Senegal. On the contrary, migration is often driven by better opportunities elsewhere rather than by livelihood stressors in the home area. Particularly the migration of young people is often an individual rather than a household decision and influenced by individual aspirations, such as aspirations for consumer goods or a better future, rather than by environmental stress.
This thesis claims that research should consider people’s capabilities to migrate or to stay as well as their individual aspirations and preferences - in addition to the household’s needs and the opportunities elsewhere. This is important in order to explain why some people stay in and others migrate from an area affected by environmental stress, though living under similar conditions. Depending on people’s capabilities to choose freely between staying and migrating and their preferences and aspirations for one or the other activity, people can either be “voluntary migrants”, “voluntary non-migrants”, “forced migrants” or “trapped people”.
Moreover, it is important to consider social trends and transformation processes in the analysis of the linkages between environment change and migration. Higher education levels and aspirations to a “modern” lifestyle among young people, for instance, might decrease the impact of environmental factors on migration, despite worsening environmental conditions.
The small leucine-rich proteoglycan biglycan (Bgn) is a part of the extracellular matrix providing structure and enhancing fibril stability. In its soluble form, biglycan is able to bind and signal via the innate immune receptors Toll-like receptor (TLR) 2 and 4, thereby activating MAP-kinases and the NF-κB pathway. In macrophages soluble biglycan induces the secretion of several cytokines and chemokines, including TNF-α, CCL2, CXCL5 and CXCL13. A unique feature of biglycan is its ability to stimulate the secretion of mature IL-1β. By orchestrating TLR2 and 4 with the purinergic P2X4 and P2X7 receptor signalling biglycan triggers the activation of the NLRP3/ASC inflammasome, which in turn activates caspase-1 to cleave pro-IL-1β to mature IL-1β. Furthermore, in several inflammatory diseases an upregulated biglycan expression is found. Enhanced levels of biglycan could be measured in plasma and inflamed tissue. In mouse models of sepsis, lupus nephritis and renal ischemic reperfusion injury, biglycan-deficiency improved the disease outcome. Overexpression of soluble biglycan on the other hand increased immune cell infiltration into the kidney by inducing cytokine and chemokine expression in a TLR2/4-dependent manner. These studies emphasise its importance in inflammatory processes, especially in the kidney. Furthermore, the pro-inflammatory effects on macrophages and diseases established biglycan as a danger signalling molecule, yet its role as a soluble molecule in plasma was not further investigated.
Although an increase of soluble biglycan in the circulation could be seen in several inflammatory diseases, the source is not fully unravelled. Previously it could be shown that macrophages and dendritic cells secrete soluble biglycan after stimulation with IL-6 and TGF-β1. However, since these cell are resident in organs and do not circulate in the blood stream their contribution to soluble biglycan levels in plasma is likely minor. Therefore, monocytes as precursor of both macrophages and dendritic cells were investigated as a possible source of circulating biglycan. Analysis of blood from septic patients revealed elevated soluble biglycan levels as well as an increased number of monocytes. Isolated monocytes from healthy volunteers incubated with the inflammatory cytokines IL-1β, IL-6 and TGF-β1 displayed increased biglycan mRNA expression and secretion of soluble biglycan into the supernatant, revealing monocytes as a producer of soluble biglycan in blood. Therefore this work was directed to further investigate the influence of soluble biglycan on circulating monocytes, with regard to sepsis.
Monocytes can be classified into three subtypes, while the classical monocytes express CD14 (CD14++CD16low), intermediate monocytes express both CD14 and CD16 (CD14++CD16+) and non-classical monocytes express mainly CD16 (CD14lowCD16++). The intermediate and non-classical monocytes make up about 10 % of all monocytes and are referred to as CD16-positive subtypes. The CD16-positive monocytes express higher levels of TNF-α and IL-1β upon stimulation and display different migration behaviour. In most inflammatory diseases an expansion of CD16-positive monocytes is observed, especially an increased number of intermediate monocytes frequently correlate with disease severity and mortality. Since septic patients had increased circulating biglycan levels and augmented CD16-positive monocytes, a possible correlation between these two parameters was investigated. Using FACS analysis of biglycan-stimulated monocytes from healthy donors revealed a significant shift from classical to intermediate and non-classical monocytes. This shift was mediated by increased expression of CD14 and CD16 on mRNA and protein levels upon biglycan treatment. Furthermore, biglycan induced the mRNA expression of the adhesion molecules ICAM-1, VCAM-1 and ELAM-1 in CD14-positive monocytes. Four hours after biglycan stimulation an increased ICAM-1 protein expression on the cell surface of classical and intermediate monocytes was observed. Additionally, biglycan-treated CD14-positive monocytes rolled and attached to pre-stimulated endothelial cells to a greater extent compared to untreated monocytes. This demonstrates that biglycan not only triggers the expression of CD14 and CD16 but also induces a functional shift of monocytes. ...
Die Parkinson Erkrankung ist die zweithäufigste neurodegenerative Erkrankung in industrialisierten Ländern. Die charakteristischen Symptome sind schwere Beeinträchtigungen des Bewegungsablaufes welche auf den Verlust dopaminerger Neurone der Substantia nigra und der damit einhergehenden Reduktion des striatalen Dopamin Gehaltes zurückzuführen sind. Alpha-Synuklein (SNCA) ist ein Protein welches zum einen mit sporadischen aber auch mit idiopathischen Erkrankungen assoziiert ist. Mutationen welche einen Funktionsgewinn von SNCA zur Folge haben konnten mit autosomal dominanten Varianten der Parkinson Erkrankung assoziiert werden und genetische Veränderungen an beiden Genenden agieren als Risikofaktor für sporadische Formen der Erkrankung. Des Weiteren wird SNCA als Hauptbestandteil der Lewy Körperchen gefunden, einem pathologischen Kennzeichen der parkinsonschen Erkrankung. Die charakteristischen Bewegungsstörungen können mittels L-DOPA, einer metabolischen Vorstufe von Dopamin, behandelt werden. Neben dem enorm positiven Effekt auf die Bewegungsstörungen, geht die Behandlung mit L-DOPA jedoch auch mit ernsten Nebenwirkungen einher, welche als Levodopa induzierte Dyskinesien (LID) beschrieben werden.
Ziel der Arbeit war die Analyse von Effekten eines SNCA Funktionsgewinns sowie des Pink1 Funktionsverlustes auf molekulare Signalwege der synaptischen Plastizität unter Verwendung dreier PD Mausmodelle (A53T-SNCA überexprimierendes Modell (PrPmtA), Pink1KO Modell sowie A53T-SNCA + Pink1KO Doppelmutante (DM)). Es wurden Kandidatengene welche eine Rolle für synaptische Plastizität spielen in 6 Monate alten Mäusen aller drei PD Mauslinien untersucht. Die Analyse von PrPmtA zeigte erhöhte mRNA Spiegel von Glutamatrezeptor-Untereinheiten und von Kandidatengenen welche eine Rolle bei der synaptischen Signalweiterleitung spielen, sowie reduzierte mRNA Spiegel von IEGs und Transkriptionsfaktoren. Die Analyse der DM zeigte nur geringe Expressionsänderungen der Glutamatrezeptor-Untereinheiten und die Analyse von IEGs und Transkriptionsfaktoren zeigte erneut reduziert mRNA Spiege. In Pink1KO Tieren konnten nur minimale Expressionsveränderungen der Kandidatengene gefunden werden, was den Schluss zulässt, dass die zuvor beschriebenen Expressionsveränderungen in PrPmtA und DM Mäusen eindeutig auf den SNCA Funktionsgewinn zurückzuführen sind. Um frühe Effekte des SNCA Funktionsgewinns zu studieren wurde die Analyse auf 3 Monate alte PrPmtA Mäuse ausgeweitet. Diese ergab, Expressionsveränderungen für Homer1, cFos, NOR1, Nurr1 und Nur77.
In einem weiteren Versuchsansatz wurde die Auswirkung des SNCA Funktionsgewinns auf das Verhalten sowie auf molekulare Parameter nach Apomorphin Behandlung analysiert. Die Analyse ergab ein erhöhtes Niveau an unwillkürlichen Bewegungsmustern mit stereotypen und dystonischen Eigenschaften in PrPmtA im Vergleich zu Wildtypen (wt). Die molekulare Analyse von striatalem Gewebe wurde zu zwei Zeitpunkten durchgeführt, 30 min nach Apomorphin Injektion und 100 min nach Injektion. Die Analyse von striatalem Gewebe welches zum Zeitpunkt 30 min nach Injektion entnommen wurde ergab eine erhöhte Apomorphin abhängige Phosphorylierung von ERK1/2, sowie eine erhöhte Apomorphin abhängige Expression von Dusp1, Dusp6 und cFos in transgenen und wt Tieren. Genotyp abhängige Unterschiede ergaben sich für cFos, welches signifikant höher in PrPmtA induziert wurde. 100 min nach Apomorphin Injektion ergab die gleiche Analyse eine erhöhte Apomorphin abhängige Phosphorylierung von ERK1 und eine erhöhte Apomorphin abhängige Expression von Dusp1, Dusp6, cFos und Nur77 in PrPmtA im Vergleich zu wt. Die Daten unterstreichen die fundamentale Rolle von SNCA auf die Neurotransmission und synaptische Plastizität und zeigen auf, dass PrPmtA ein zuverlässiges Modell für die Analyse von präsynaptischer Dysfunktion in Frühstadien der Parkinson Erkrankung darstellt.
Der letzte Versuchsansatz stellt die Charakterisierung des DM-Mausmodells welches sich durch einen starken Phänotyp auszeichnet, sowie die Analyse des Pink1 Effektes auf die SNCA induzierte Neurotoxizität dar. DM-Tiere zeigen deutlich reduzierte Spontanmotorik im Alter von 3 Monaten sowie einer progressiven Lähmung der Hinterläufe, was Anlass zu einer immunhistologischen Charakterisierung mittels Schnitten des Gehirns und Rückenmarks gab. Die histologische Analyse zeigte pSer129-SNCA, p62/SQSTM1 und Ubiquitin positive Aggregate innerhalb der grauen Substanz des Rückenmarks sowie innerhalb einer neuronalen Zellpopulation welche dorsal der Substantia nigra angeordnet ist. Das histologische Erscheinungsbild wurde spezifisch in gelähmten DM-Tieren gefunden und nicht in Einzelmutanten oder DM-Tieren ohne Lähmung. Dieses Modell stellt ein wertvolles Instrument für die Identifizierung von pathologischen Mechanismen und Signalkaskaden welche beiden Parkinson relevanten Genen gemeinsam sind, dar.
In the present work, mismatch negativity (MMN) was used to examine the contribution of spectral vs. temporal perceptual features to vowel length discrimination in children and adults. Three age groups (adults vs. 9-10 years vs. 10-11 years olds) have been taken to examine developmental effects on vowel length perception. Natural (i.e., spectrotemporal) vowel length differences were compared with (artificially modified) stimulus pairs varying only in temporal or spectral characteristics to contrast spectral, temporal and spectrotemporal processing.
The result indicates that, while adults integrate spectral and temporal aspects of the speech signal in an additive way, children of 9-10 years of age sequentially process both features. However, vowel length processing is found to become adultlike at the age of 10-11 years.
Im Rahmen dieser Arbeit werden verschiedene Modellsysteme untersucht, die Metriken der klassischen Allgemeinen Relativitätstheorie mit Erweiterungen vergleichen, in denen Ereignishorizonte nicht existieren müssen. Die untersuchten Korrekturterme sind durch Schwachfeldmessungen, wie sie zum Beispiel in unserem Sonnensystem durchgeführt werden, nicht überprüfbar. Es ist deshalb nötig solche Systeme zu betrachten, in denen die vollständigen Gleichungen berücksichtigt werden müssen und keine Entwicklungen für schwache Felder gemacht werden können. Es gibt eine Reihe von astrophysikalischen Systemen, die diese Bedingungen erfüllen, wie das Galaktische Zentrum oder Doppelsternsysteme.
Im zweiten Kapitel der Arbeit werden Testteilchenorbits in einem Zentralpotential beschrieben und Unterschiede zwischen der klassischen und einer modifizierten Kerr-Metrik herausgearbeitet. Drei neue Phänomene der modifizierten Metrik gegenüber der Klassischen treten hier in Erscheinung. Zum einen haben Teilchen, die sich auf prograden Bahnen um den Zentralkörper drehen, ein Maximum in ihrer Winkelgeschwindigkeit. Zum anderen ist das Phänomen des frame-draggings deutlich schwächer ausgeprägt. Schließlich tritt ein letzter stabiler Orbit für entsprechend schnell rotierende Zentralkörper nicht mehr auf. Gleichzeitig sind die Unterschiede in den beiden Metriken für große Abstände (r > 10m) nahezu vernachlässigbar. In Kapitel 3 werden diese Ergebnisse auf zwei unterschiedliche Modelle zur Beschreibung von Akkretionsscheiben angewendet. Untersucht wird zum einen das Verhalten der Eisen-Kα-Emissionslinie und zum anderen der Energiefluss aus einer Akkretionsscheibe.
In der Form der Eisen-Kα-Emissionslinie gibt es eine deutliche Zunahme des rotverschobenen Anteils der Strahlung in der modifizierten Kerr-Metrik gegenüber der klassischen Kerr-Metrik. Die Akkretionsscheibe nach Page und Thorne zeigt unter Verwendung der modifizierten Kerr-Metrik eine signifikante Erhöhung der abgestrahlten Energie, wenn der Zentralkörper so schnell rotiert, dass kein letzter stabiler Orbit mehr auftritt. Zusätzlich gibt es hier in der Scheibe einen dunklen Ring im Vergleich zu den Bildern höherer Ordnung, die in der klassischen Kerr-Metrik auftreten. Erklärbar sind diese Phänomene dadurch, dass sich Teilchen auf stabilen Bahnen in der modifizierten Kerr-Metrik näher an den Zentralkörper heran bewegen können, als es in der klassischen Kerr-Metrik der Fall ist. Die Rotverschiebung ist für beide Fälle annäherend gleich.
Kapitel 4 gibt eine kurze Einführung in die Beschreibung von Gravitationswellen im Rahmen der linearisierten Allgemeinen Relativitätstheorie. Hier wird als Modell ein Binärsystem, wie etwa der Hulse-Taylor-Pulsar, betrachtet. Die Unterschiede zwischen der klassischen Theorie und einer Beschreibung unter Hinzunahme von Zusatztermen sind hier erwartungsgemäß sehr gering, da die Linearisierung der Gleichungen dazu führt, dass Starkfeldeffekte vernachlässigt werden. Für große Abstände, was in diesem Fall auch schwache Felder impliziert, sind die Erweiterungen der Gleichungen vernachlässigbar. Hier werden zum Teil auch Effekte in der klassischen ART vernachlässigt.
In Kapitel 5 befindet sich ein kurzer Ausblick in die 3+1-Formulierung der Einsteingleichungen für die numerische Beschreibung von Gravitationsphänomenen. Diese Beschreibung ermöglicht es auch komplexe Systeme ohne viele nähernde Annahmen genau beschreiben zu können. Diese Systeme können zum einen Akkretionsscheiben um kompakte Objekte sein, aber auch die Verschmelzung von zwei massiven Objekten und die damit verbundenen Gravitationswellensignale. Dadurch lassen sich die Vorhersagen der ART oder etwaiger Erweiterungen präziser modellieren.
Die vorgestellten Ergebnisse liegen innerhalb der Einschränkungen durch aktuelle Messungen. Zukünftige Messungen wie genauere Beobachtungen des Galaktischen Zentrums durch das Event Horizon Telescope sind aber voraussichtlich dazu in der Lage zwischen den untersuchten Metriken zu unterscheiden.
Termites are important ecosystem engineers of the savanna biome, with the large mounds of fungus-cultivating termites being sources of habitat heterogeneity and structural complexity in African savanna landscapes. Studies from different localities throughout Africa have shown that termite mounds have a strong influence of diversity and composition of plant communities. However, most research has been conducted only at the local scale, and integrating knowledge across Africa is hampered by different methodology of studies and differing environmental context. Little is known about the variation in vegetation composition on termite mounds compared to the surrounding savanna at the regional scale and at the landscape scale, and the main determinants of plant communities on mounds are yet to be ascertained.
This thesis aimes at better understanding the influence of termite mounds on vegetation compared to the surrounding savanna across spatial scales. Three research projects analyse vegetation data and soil data from paired mound and savanna plots in West Africa. The first project examines the influence of termite-induced heterogeneity on plant diversity and vegetation composition at a regional scale, following a bioclimatic gradient from the Sahel of Burkina Faso to the Sudanian vegetation zone in North Benin. The second Project analysed variation of vegetation on and off mounds at the landscape scale in Pendjari National Park, North Benin. The third is a monitoring study over the course of two years, exploring dynamics of juvenile woody plant communities on mounds and in the surrounding savanna at a local scale. The thesis thus provides the first comparative quantitative analysis across scales of mound and savanna vegetation and the drivers of the mound–savanna difference in vegetation.
Synthesizing across scales, its results confirm that termite mounds strongly contribute to savanna plant diversity, even though mounds are not generally more species rich than the surrounding savanna. Variation in mound vegetation is much higher along climatic and soil gradients than previously acknowledged. Mound vegetation differs from the surrounding savanna in the whole study area and in each sampled savanna type, with the strongest differences occurring at the most humid study sites. A large proportion of the differences between mound and savanna vegetation is explained by clay enrichment and related soil factors, such as cation concentrations. Plants on mounds thus benefit from favourable soil conditions, including higher fertility and higher water availability, which is also mirrored by the higher abundance and basal area of juvenile woody plants found on mounds. The variation in mound vegetation between study sites across scales results in part from local differences in soil composition and from climatic differences that influence the regional distribution of species. Different sets of characteristic mound species are identified in each project. Specific plant families and traits like succulency, lianescence, and adaptations to zoochory are found to be overrepresented in mound communities.
In addition to the findings in this thesis, remaining parts of the variation in mound vegetation between study sites could likely be explained by investigating further factors. Specifically, mound vegetation depends on habitat context, which includes available species pools, spatial distribution of mounds, biotic interactions with dispersers and herbivores, fire, and also anthropogenic influence. The high proportion of species with adaptations to zoochory found on mounds, for example, indicates that animal dispersers should be of particular importance for vegetation on termite mounds. Herbivory and fire regime, which are known to contribute to the diversity and community composition of the mound–savanna system, also show strong local variation, not least because of anthropogenic influence.
In conclusion, termite mounds play a crucial role in maintaining heterogeneity and plant diversity in the savanna across scales. Ecosystem services provided by termites, especially considering long-term effects on soil fertility and ecosystem resilience, are most likely undervalued. Mounds should be considered in management plans from local to regional, transnational scales as a matter of course, accompanied by further research on the role of termite mounds in savanna ecology on a longer temporal scale. The research presented here thus provides a basis for future studies on termite mound vegetation that should specifically consider the biotic and abiotic context of the mound–savanna system.
Tectonin β-propeller containing protein 2 (TECPR2) was first identified in a mass- spectrometric approach as an interactor of GABARAP, an ATG8-family protein playing a role in autophagy. The mammalian ATG8 protein family consists of seven members, namely MAP1LC3A (LC3A), MAP1LC3B (LC3B), MAP1LC3C (LC3C), GABARAP, GABARAPL1 and GABARAPL2. All share an ubiquitin-like core and possess two additional N-terminal α-helices, which are important for the distinct functions of the proteins. First determined in various organelles the ATG8 proteins are shown to be involved in autophagy, supporting the formation and cargo recruitment of autophagosomes, the vesicles transporting cargo for autophagic degradation.
Autophagy is the process of recycling cytoplasmic contents by degradation of misfolded proteins or damaged organelles in order to supply nutrients. Also clearance of pathogens can be achieved via autophagy. Importantly, LC3B is incorporated into the autophagosomal membrane and is therefore used as the main marker for autophagosomes. Previous studies exhibited that depletion of TECPR2 leads to a loss of LC3B-positive structures in cells, which suggests TECPR2 to positively regulate autophagic processes.
A frame shift deletion in the gene encoding for TECPR2 causes the generation of a premature stop codon and subsequent an unstable version of the protein, which is then degraded. Mutation in the TECPR2 gene triggers a neurodegenerative disorder termed hereditary spastic paraparesis (HSP). HSPs are a diverse group of neurodegenerative diseases that are characterized by spasticity in prevalent lower extremities and were mediated by a loss of axonal integrity of the corticospinal motor neurons. In the context of HSP more than 50 gene loci were identified by now. While TECPR2 is a human ATG8 binding protein and positive regulator of autophagy causing a form of HSP, the exact function of TECPR2 is unknown.
This study primarily focused on the determination of TECPR2’s binding mode to ATG8 proteins in vitro and in cells. The association of TECPR2 to all ATG8-family proteins was confirmed in in vitro pulldown experiments. Following fragment-based binding and peptide array experiments, the LC3-interacting region (LIR) of TECPR2 could be verified with mutants of TECPR2 lacking the LIR motif. Nuclear magnetic resonance (NMR) and isothermal titration calorimetry (ITC) were conducted to gain deeper insights into the binding preference to the different ATG8-family members. Moreover, the crystal structure of TECPR2-LIR was solved. In cells colocalization studies with overexpressed ATG8 proteins unraveled a preferential binding to the LC3-subfamily.
Further, mass spectrometric analysis revealed novel association partners of TECPR2: SEC24D, HOPS and BLOC-1, all of those participating in different endomembrane trafficking pathways. Interaction and colocalization of TECPR2 with these components was validated with several immunoprecipitation experiments and the N-terminal part of the protein comprising the WD40-domain could be defined as the binding site for all three of the association partners. In further approaches, the requirement of the LIR-motif and the necessity of the availability of LC3 protein for the particular interactions were determined. Interestingly, in the absence of LC3C the binding of TECPR2 to SEC24D was completely disrupted whereas a loss of LC3B only resulted in a decreased association. Notably, the binding proteins were not subjected to autophagosomal degradation, indicating that TECPR2 may operate as a multifunctional scaffold protein. While depletion of TECPR2 destabilized HOPS and BLOC-1, the autophagy defect observed in TECRP2-deficient cells could not be attributed to functional impairment of these two complexes.
Moreover, loss of TECPR2 led to a decline in protein levels of SEC24D and of its heterodimer partner SEC23A. Thus, TECPR2 is required to regulate the protein levels of SEC23A and SEC24D and subsequently the formation of the heterodimers. Together, SEC24D and SEC23A form the inner coat of COPII vesicles. These vesicles are responsible for the anterograde transport of cargo from the ER toward the Golgi compartment. COPII-coated vesicles are secreted form ER at distinct sites, termed ER exit sites (ERES). The small GTPase SAR1A maintains the vesicle budding, coating and secretion at the ERES. Together with SEC13, SEC31 forms the outer coat of the COPII vesicles and therefore serves as a general ERES marker.
Consistent with a defect in COPII coat assembly, the number of ERES diminished in the absence of TECPR2. These phenotypes could be rescued by the wildtype TECPR2 protein but not by the LIR-mutant. Intriguingly, these results were mimicked by depletion of LC3C, which localized to ERES. By monitoring the release of various cargos from ER in dependency of TECPR2 or LC3C, a role of both proteins in ER export was determined. These facts indicated that TECPR2 cooperates with LC3C to facilitate COPII assembly, ERES maintenance and ER export. Notably, fibroblast derived from a HSP patient carrying mutated TECPR2 showed diminished SEC24D protein levels and delayed ER export.
Concurrent with emerging evidence for a role of ERES in autophagosome formation, depletion of TECPR2 or LC3C or overexpression of a constitutive inactive SAR1 mutant reduced puncta formation of the early autophagosomal protein WIPI2.
In summary, this study uncovered a role for TECPR2 in ER export at ERES through interaction and stabilization of SEC24D, a COPII coat protein. This process also depended on ATG8-family protein LC3C, which is localized at ERES. Both proteins are required for correct COPII-mediated secretion. Moreover, the presence of TECPR2 and LC3C on ER allows development of omegasomes, membranous structures budding ER to form autophagosomes, by stabilization of WIPI2 and therefore contribute to autophagosome formation.
Die größte Gruppe der Krebserkrankungen bei Kindern sind die Leukämien. Die größte Untergruppe stellen dabei die Leukämien unter Beteiligung des MLL-Gens auf Chromosom 11q23 dar. Die bei MLL-Translokationen gefundenen Partnergene sind äußerst vielfältig. Der häufigste Partner ist jedoch das AF4-Gen auf Chromosom 4. Die bei der t(4;11)-Translokation entstehenden Fusionsproteine MLL-AF4 und AF4-MLL sind die Auslöser der Leukämie, wobei in unterschiedlichen Forschungsarbeiten beiden Fusionsproteinen eine Transformatorische Wirkung bescheinigt werden konnte. Das Wildtyp MLL-Protein liegt in der Zelle in einem Multiproteinkomplex vor, der durch seine Histon-Methyltransferase-Aktivität an Lysin 4 des Histon H3, zu einer offenen Chromatinstruktur führt und dadurch für die Transkriptionsinitiierung essentiell ist. Eine besondere Rolle kommt den MLL-Protein bei der Aufrechterhaltung von epigenetischen Signaturen beispielsweise bei der Embryogenese oder dem Durchlaufen des Zellzyklus zu. Im Gegensatz dazu nimmt der F4-Multiproteinkomplex eine wichtige Stellung in der Transkriptionselongation ein und ermöglicht der RNA-Polymerase II zusammen mit seinen Komplexpartnern die Elongation der mRNA. Die Taspase1 wurde als das Protein entdeckt, dass für die Prozessierung des MLL-Proteins verantwortlich ist und es an den Schnittstellen CS1 bzw. CS2 proteolytisch spaltet. Die hierbei entstehenden Fragmente N320 und C180 können daraufhin dimerisieren und werden gegenüber einem proteasomalen Abbau stabilisiert. Diese Taspase1-Schnittstellen sind auch in dem Fusionsprotein AF4-MLL enthalten und ermöglichen die Stabilisierung des AF4-MLLs nach proteolytischer Spaltung gegenüber seinem proteasomalen Abbau. Die Taspase1 ist eine Threonin-Aspartase aus der Familie der Typ-2 Asparaginasen zu deren weiteren Vertretern die L-Asparaginase sowie die Glycosylasparaginase zählen. Als einziger Vertreter dieser Gruppe ist die Taspase1 jedoch eine Protease. Gemein ist allen Vertretern der Familie, die autoproteolytische Aktivierung des exprimierten Proenzyms hin zu einer katalytisch aktiven Form. Im Falle der Taspase1 kommt es dabei zu einer Spaltung in die als Heterodimer vorliegenden α- und β-Untereinheiten. Das katalytische Nukleophil der aktiven Taspase1 ist dabei das N-terminale Thr234 der β-Untereinheit. Neben ihrer Rolle als Protease des AF4-MLLs und der damit verbundenen Stabilisierung des Onkogens, wird der Taspase1 auch bei den soliden Tumoren eine Rolle als für die Transformation wichtiges Protein zugewiesen, was sich in ihrer häufigen Überexpression in verschiedenen Tumorarten widerspiegelt. Die Taspase1 ist demnach ein interessantes Ziel für die Wirkstoffentwicklung. Hierfür ist die Generierung eines quantitativen Aktivitätstest besonders wichtig und war Ziel dieser Arbeit. Der entwickelte Aktivitätstest basiert auf einem Reporter bestehend aus den beiden Fluoreszenzproteinen TagBFP sowie TagGFP2, die über eine Taspase1 Schnittstelle verbunden wurden. Dieses FRET-Paar lässt dabei die kontinuierliche Beobachtung der Reaktion zu und ermöglicht eine Auswertung der kinetischen Parameter der Reaktion. Von den beiden FRET-Reportern mit den unterschiedlichen Schnittstellen CS1 und CS2,konnte nur der mit der CS2 Schnittstelle exprimiert werden. Dieser Reporter konnte dann für die Validierung des Aktivitätstests eingesetzt werden und ließ die Bestimmung der kinetischen Parameter der Taspase1 für diese Reaktion zu. Die erhaltenen Parameter bewegen sich in einer ähnlichen Größenordnung wie schon publizierte kinetische Parameter eines Peptid-basierten Aktivitätstests. Ausgehend hiervon wurden die dnTaspase, eine dominant negative Taspase1-Mutante, kinetisch untersucht und ihre Inhibition der Taspase1-Aktivität beschrieben. Der Aktivitätstest ließ die Bestätigung der Konzentrationsabhängigen Inhibition der Taspase1-Aktivität durch die dnTaspase zu. Diese Beobachtung konnte auch in einer Zell-basierten Variante des Aktivitätstests bestätigt werden und zeigte sich ferner auch in einer Wachstumsverlangsamenden Wirkung der dnTaspase in der t(4;11)-Zelllinie SEM, wobei dieser Effekt nicht auf einer Zunahme der Apoptose der Zellen zurückzuführen war. Desweiteren wurde der Aktivitätstest benutzt, um eine Reihe von gegen das aktive Zentrum der Taspase1 gerichtete Substanzen auf ihre inhibitorische Wirksamkeit zu untersuchen.Hierbei gelang es einen Kandidat als Leitsubstanz zu identifizieren, der eine konzentrationsabhängige Inhibition der Taspase1 zeigte. Eine Bindung dieser Substanz an die Taspase1 konnte jedoch durch einen Thermal Shift Assay nicht nachgewiesen werden und ein Einfluss auf die Viabilität von proB-ALL Zelllinien konnte nicht beobachtet werden.
Künstliche Ribonucleasen, die sequenzspezifisch und effizient die Spaltung von RNA-Phosphordiesterbindungen katalysieren, könnten potenziell nicht nur als biochemische Werkzeuge dienen, sondern auch als Wirkstoffe gegen eine Vielzahl von Erkrankungen, bei denen mRNA oder miRNA involviert sind, eine wichtige Rolle spielen. Obwohl in den letzten beiden Jahrzehnten zahlreiche sequenzspezifische RNA-Spalter entwickelt wurden, bleibt die Spaltaktivität dieser Verbindungen nach wie vor deutlich hinter der ihrer natürlichen Äquivalente zurück. Die Optimierung künstlicher Ribonucleasen und grundlegend dafür die Erforschung der Faktoren, die die Spaltaktivität einer Verbindung beeinflussen, sind daher weiterhin von großem Interesse. Zwar enthalten die meisten künstlichen Ribonucleasen Metallionen, doch sind auch metallfreie RNA-Spalter, zum Beispiel auf der Basis heterocyclischer Guanidine, bekannt. Prinzipiell kann die Hydrolyse des RNA-Rückgrates durch Deprotonierung der nucleophil am Phosphoratom angreifenden 2‘-OH-Gruppe, durch Protonierung der als Abgangsgruppe fungierenden 5‘-OH-Gruppe sowie durch Stabilisierung des bei der Spaltung durchlaufenen dianionischen Phosphorans katalysiert werden. Daher sollten potenzielle RNA-Spalter in der Lage sein, sowohl als Base als auch als Säure wirken zu können, was bei einem pKa-Wert im Bereich von 7 am besten gegeben ist. Fungiert ein und dasselbe Molekül als Protonenakzeptor und -donor, so kommt es im Fall von Guanidinanaloga zu einer Tautomerisierung vom Amino- zum Iminoisomer. Eine möglichst kleine Energiedifferenz zwischen beiden Formen sollte sich daher positiv auf die Spaltaktivität auswirken. In der vorliegenden Arbeit wurde eine Reihe heterocyclischer Guanidine synthetisiert, deren pKa-Werte bestimmt und die jeweiligen Energiedifferenzen zwischen Amino- und Iminotautomer grob mittels AM1-Rechnungen abgeschätzt. In Spaltexperimenten wurden Cy5-markierte RNA-Substrate mit den verschiedenen Verbindungen inkubiert (Spalter-Konzentration: 2 bzw. 10 mM). Die Analyse und Quantifizierung der Spaltprodukte erfolgten anschließend mithilfe eines DNA-Sequenziergerätes. Alle untersuchten und ausreichend löslichen Substanzen, die sowohl einen geeigneten pKa-Wert (6 – 8) als auch eine niedrige Energiedifferenz zwischen Amino- und Iminotautomer (≤ 5 kcal/mol) aufwiesen bzw. bei denen nur der pKa-Wert oder nur die Energiedifferenz in geringem Maße vom Idealwert abwich, spalteten RNA, wenn auch teilweise nur mit einer geringen Aktivität. In den Spaltexperimenten erwiesen sich Guanidinanaloga mit einem großen aromatischen System als besonders aktiv, allen voran 2-Aminoperimidin und seine Derivate, die auch bei Konzentrationen unter 50 µM Spaltaktivität zeigten. Gleichzeitig offenbarten diese Verbindungen in Fluoreszenzkorrelationsspektroskopie Experimenten eine große Tendenz zur Aggregation mit RNA, so dass die Spaltung in diesen Fällen möglicherweise nicht durch Einzelmoleküle, sondern durch Aggregate erfolgte. Um RNA-Substrate auch sequenzspezifisch spalten zu können, wurden PNA-Konjugate des bereits bekannten RNA-Spalters Tris(2-aminobenzimidazol) hergestellt, wobei der Spalter über eine neue, quecksilberfreie Route synthetisiert wurde. Es konnte gezeigt werden, dass diese PNA-Konjugate RNA sequenzspezifisch mit einer Halbwertszeit von etwa 11 h spalten, was im Rahmen der Halbwertszeit vergleichbarer DNA-Konjugate liegt. Um zu untersuchen, ob 2-Aminoperimidine auch als Einzelverbindungen aktiv sind, wurden zwei PNA-Konjugate von am Naphthylring substituierten 2-Aminoperimidin-Derivaten synthetisiert. In addition to infectious viral particles, hepatitis B virus-replicating cells secrete high amounts of SVPs, which are ssembled by HBsAg in the shape of spheres and filaments but lack any capsid and genome. Filaments are characterized by a much higher amount of the surface protein LHBs as compared to spheres. Spheres are
released via the constitutive secretory pathway, while viral particles are ESCRT-dependently released via MVBs. The interaction of virions with the ESCRT machinery is mediated by α-taxilin that connects the PreS1 domain of LHBs with the ESCRT-component tsg101. Since viral particles and filaments contain a significant amount of LHBs, it is unclear whether filaments are secreted as spheres or released like viral particles. To study the release pathways of HBV filaments in the absence of viral particles, A core-deficient
HBV mutant (1.2×HBVΔCore) was generated by site-directed mutagenesis based on wt1.2x HBV. The start codon of core protein was mutated into stop codon, which was confirmed by DNA sequencing. Data from HBsAg ELISA, Western blot, immunofluorescence microscopy and immunoelectron microscopy showed that the lack of core protein did neither affect the production nor the secretion of HBV SVPs. The intracellular distribution of
LHBs and SHBs showed no difference between wtHBV and the core-deficient mutant expressing cells. Therefore, this system is suitable to investigate the release pathway of HBV filaments in the absence of viral particles. Confocal microscopy analysis of cells cotransfected core-deficient mutants with peYFPRab7 as marker for the endosomal/MVB pathway or with pGalT-eGFP as marker for the trans Golgi apparatus showed that YFP-Rab7, but not GalT-GFP, partially colocalized with LHBs. Furthermore, LHBs could be found in dilated MVBs by immune electron microscopy of ultrathin sections. This was confirmed by isolation of MVBs by cell fractionation using discontinuous sucrose gradient ultracentrifugation and percoll-based linear gradient ultracentrifugation, indicating that filaments enter MVBs in the absence of virion formation. Moreover, inhibition of MVB biogenesis by the small molecular inhibitor U18666A significantly abolished the release of filaments in a dose-dependent manner, but no inhibition could be observed in the production. In contrast, no inhibition on the secretion and production of spheres could be
detected. Inhibition of ESCRT-functionality by coexpression of transdominant negative mutants (Vps4A, Vps4B, CHMP3) abolished the release of filaments while secretion of spheres was not affected. These data indicate that in contrast Abstract 73 to spheres while are secreted via the secretory pathway, filaments are released via ESCRT/MVB pathway like infectious viral particles.
In order to investigate the diversity of the western honeybee, Apis mellifera L., in West and Central Africa, a total of 204 colonies were sampled from 44 localities in four countries – Nigeria, Niger, Cameroon and Chad. 86 of these colonies, from 23 localities, were subjected to full morphometric analysis. In a principal component analysis (PCA) of the morphometric data, the colonies formed a single cluster. It also revealed that overall size of the body was the most important source of variation between the colonies. A hierarchical structure analysis, followed by a stepwise discriminant analysis, classified the colonies into three distinct morphoclusters; however, these clusters were not geographically demarcated. In another PCA carried out with the samples under investigation and reference samples of A. m. adansonii, A. m. jemenitica and A. m. scutellata, the colonies under investigation again formed one cluster which lying over and extended beyond the clusters of the reference subspecies. This is suggestive of a wider variation in size in the bees under investigation. In a stepwise DA, 94.2% of cross-validated grouped cases were correctly classified and the distances between group centroids were highly significant (p < 0.0005) according to F-statistic. 61 and 22 of the 83 colonies under investigation were assigned to A. m. jemenitica and A. m. adansonii, respectively. Mitochondrial DNA analysis was carried out on 148 colonies from 39 localities. Four mitochondrial haplotypes, previously reported from Africa and belonging to the African mitochondrial lineage, A, were detected: A1 (n = 62), A4 (n = 70), A4' (n = 15) and A14 (n = 1). The overall haplotype diversity was low (h = 0.478 ± S. E. 0.057). A chi-square test for association was conducted between haplotypes and type of vegetation, latitude, longitude, altitude, temperature and rainfall, severally. There was a statistically significant association between haplotype and each of the six variables and the association was strong with latitude, moderate with vegetation and rainfall and weak with the remaining variables. The neighbour-joining, maximum likelihood and maximum parsimony trees, obtained from sequence variation of the cytochrome b gene of mitochondrial DNA, showed that the samples, from the current study, unambiguously clustered with the reference sequences of A. m. scutellata from Kenya, but without showing further subdivision within this sub-Saharan cluster. 133 workers (one per colony) collected from 38 localities were subjected to microsatellite analysis. A total of 292 different alleles were recorded for the 15 microsatellite loci used. All microsatellite loci were polymorphic and the number of different alleles per locus ranged between 10, in locus At163, and 31, in locus A029. Heterozygosity (or gene diversity) was high in all loci. The unbiased expected heterozygosity, which is a better expression of gene diversity, was 0.861 ± S.E. 0.017. The overall FST value, which is a good estimate of genetic differentiation of populations, was very low: 0.007 ± S.E. 0.001 (0.001 - 0.014). AMOVA and Bayesian assignment showed no differentiation of the investigated populations. Based on morphometric analysis, the results of this study present the honeybees of western Africa as a single entity with an internal variation which lacks a geographical demarcation. Consequently the results do not support the splitting of the honeybees of the region into the two subspecies, A. m. adansonii and A. m. jemenitica, as reported in the literature. More morphometric, molecular, physiological and behavioural studies are required to confirm the taxonomic status of the honeybees of the region. Meanwhile, the use of A. m. adansonii, as the sole sub-specific name for the honeybees of West and Central Africa, is recommended.
Die Biosynthese der Fettsäuren (FS) ist in Eukaryoten und Bakterien ein hochkonserviert zentraler Stoffwechselweg, der in zwei strukturell verschiedenen Systemen ausgeführt wird. Die meisten Bakterien, Parasiten, Pflanzen und Mitochondrien nutzen ein Fettsäuresesynthase Typ-II (FAS-II) System. Bei FAS II Systemen sind alle katalytischen Domänen separate lösliche Proteine. In Eukaryoten wie auch den Bakterien Corynebakteria, Mycobakteria, Nocardia (Klasse der CMN Bakterien) liegen die katalytischen Domänen fusioniert auf einer Polypeptidkette vor, die zu einem Multienzymkomplex der Fettsäuresynthase Typ I (FAS-I) assemblieren. Die Architektur der FAS-I zeigt große Unterschiede; die X förmige Säuger-FAS-I (Maier et al., 2006), sowie die fassartigen Enzyme der Pilz FAS-I (Jenni et al., 2007; Leibundgut et al., 2007; Lomakin et al., 2007; Johansson et al., 2008) und der bakteriellen FAS-I (Boehringer et al., 2013; Ciccarelli et al., 2013). Zwischen Pilz- und bakterieller FAS-I gibt es trotz des ähnlichen Aufbaus bedeutende Unterschiede. Mycobakterium tuberculosis, der Auslöser von Tuberkulose (TB), an der jährlich über eine Million Menschen weltweit sterben (WHO, 2014), synthetisiert durch eine Symbiose von FAS-I, FAS-II und der Polyketidsynthase-13 Mykolsäuren. Durch die Mykolsäuren ist M. tuberculosis resistent gegen äußere Einflüsse. FAS-I ist in die Synthese der Vorstufen der Mykolsäuren involviert. Sie stellt im Kampf gegen TB ein potentielles Inhibierungstarget dar.
Strukturell war die bakterielle FAS-I beim Beginn der vorliegenden Arbeit, nur durch negative-stain-Elektronenmikroskopie (EM) Aufnahmen aus dem Jahr 1982 charakterisiert (Morishima et al., 1982). In dieser Arbeit konnte die bakteriellen FAS I aus M. tuberculosis (MtFAS), sowie Corynebacterium ammoniagenes (CaFAS) und Corynebacterium efficiens (CeFAS) strukturell untersucht werden. Dies geschah mit den Methoden negative-stain-EM, Einzelmolekül-Cryo-EM (Cryo-EM), Cryo EM Tomographie (CET) und Röntgenkristallographie.
Anhand von CeFAS-Kristallen konnte erstmals durch Röntgenkristallographie die Struktur einer bakteriellen FAS-I bestimmt werden. Zudem wurde die hohe konformationelle Flexibilität der bakteriellen FAS-I mit mehreren Methoden gezeigt. Für die CaFAS konnte mit Cryo-EM initiale Prozesse der Proteinkristallbildung abgebildet werden.
Structural characterization of stressosome complexes by single-particle cryo-electron microscopy

The stressosome is a Mega Dalton macromolecular complex involved in stress adaptation in bacteria. Stressosomes are considered as stress signaling hubs. They are able to perceive a variety of different stress stimuli and transduce them into one single cellular answer, which is the initialization of a transcriptional up-regulation of hundreds of different genes encoding for universal but also very specific stress response proteins.
The stressosome of Bacillus subtilis became a prime example for this intriguing stress-triggered transcriptional regulation when its architecture was determined by Single-particle cryo-electron microscopy (cryo-EM) in 2008. In Gram-positive Bacillus species, the stressosome complex senses changes in salt concentration, ethanol content, blue-light, heat or acid stress contributing to the general stress response by activation of the alternative σB factor. σB is a transcriptional promoter that initiates the transcription of over 150 general stress genes, e.g., genes that encode osmolyte transporters to counteract osmotic and chill stress. The B. subtilis stressosome (stressosome_Bc) is composed of multiple copies of the 3 proteins: RsbR, RsbS and RsbT. These three Rsb proteins (Regulator of Sigma B) are found clustered in one operon forming the conserved RST module. RsbS and RsbR are scaffold proteins comprising a STAS domain, respectively. Because these domains are dominantly associated to sulfate transporters and anti-sigma antagonist they were named STAS domains, however, they were also identified in other sensor proteins. In the stressosome they form the internal ball-shaped core, while the N-terminal globin-fold sensor domain of RsbR, protruding to the outside, facilitates stress sensing. It is assumed that the stress signal is transduced to the stressosome core via the STAS domain resulting in conformational changes of the core. These changes affect the binding of the third protein, RsbT, a serin-threonine kinase. As a direct consequence of stress sensing the RsbT kinase is released from the complex to start an activation cascade involving the stepwise activation of RsbU, V, W, and X, which are all part of the same operon, and finally of σB. In Bacillus species, several RsbR orthologs were identified varying mainly in the sequence of the N-terminal sensor domains. It is assumed that the stressosome_Bc assembles with a still unknown combination of RsbR orthologs allowing for the broad spectrum of stress stimuli that can be processed in vivo. The pathogenic bacteria Listeria monocytogenes is a close relative of Bacillus. Its potent stress response allows Listeria to survive the harsh environmental conditions during host infection and therefore the stress regulation machinery is contributing heavily to the virulence of this pathogen. In Listeria the Rsb operon is conserved and highly homologous to the Bacillus one. In the frame of this thesis, the in vitro assembly of Listeria innocua stressosomes was shown for the first time by Single-particle (SP) negative stain EM. Moreover, binding of Listeria RsbT to the assembled RsbR-RsbS complex was demonstrated biochemically.
Despite the conservation of the RST-module the entire Rsb operon is not conserved in the bacterial kingdom suggesting that signal transduction and regulation of gene expression might occur by very different mechanisms in stressosomes of different species. We have focused here on a stressosome type from the Gram-negative pathogen Vibrio vulnificus that is quite distinct from the Bacillus ones with respect to (1) the missing conservation of the Rsb operon, (2) the role of RsbT, (3) the activation of a different transcriptional promoter, and (4) the absence of additional RsbR orthologs. Interestingly, there is only one RsbR protein encoded in the genome. This one contains a Haem-group in its N-terminal domain being oxygen sensitive. It is assumed that the Vibrio stressosome perceive only oxidative stress and that regulation occurs via a diguanylate cyclase with a GAF domain that synthesizes the second messenger c-di-GMP from GTP.
We have started a structure determination of the Vibrio vulnificus stressosome by SP cryo-EM to elucidate the differences in the molecular mechanism of stress sensing in divers stressosome types. A 3D map of the oxidized (activated) Vibrio vulnificus stressosome was determined to 7.6 Å resolution revealing an increased flexibility of both the core and the N-terminal sensor domains in comparison to the Bacillus stressosome suggesting that our structure has trapped for the first time an active state of a stressosome complex. A 3D map of the stressosome core to 7 Å resolution allowed fitting of a homology model of the Vibrio stressosome based on the Bacillus stressosome as template. The conformational changes could be attributed to the entire core, which was confirmed by MD simulations.
Rotary adenosine triphosphate (ATP)ases are ubiquitous, membrane-bound enzyme complexes involved in biological energy conversion. The first subtype, the so-called F1Fo ATP synthase, predominantly functions as an ATP synthesizing machinery in most bacteria, mitochondria and chloroplasts. The vacuolar subtype of enzyme, the V1Vo ATPase, operates as an ATP driven ion pump in eukaryotic membranes. The subtype found in archaea and some bacteria is called A1Ao ATP (synth)ase and is capable of working in both directions either to synthesize ATP or to generate an ion motive force by consuming the same.
All the three above-mentioned subtypes of rotary ATPases work as nanomolecular machines sharing a conserved mechanism to perform the energy conservation process. The simplest form of these enzymes is the bacterial F1Fo ATP synthase. Here, ions are channelled via the membrane stator subunit a to the rotor ring of the enzyme. After almost a complete rotation of the ring the ions are released again on the other side of the membrane. This rotation is further transmitted via the central stalk to the soluble part of the enzyme, the F1-complex, where conformational changes within the nucleotide binding sites result in the synthesis of ATP from ADP and Pi.
The rotor or c-ring of the enzyme is the key protein complex in mediating transmembrane ion translocation. Several structural and biochemical methods have been applied in the past years to study the rotor rings from many different organisms. The results revealed that the stoichiometry of a c-ring of a given species is constant while it can vary between different species within a range of 8 to 15 c subunits. The c-ring stoichiometry determines directly the number of ions transported through Fo per rotation whereby three molecules of ATP are concurrently synthesized in the water-soluble F1 headgroup. Hence the number of c subunits has an important influence on the bioenergetics of the corresponding enzyme and thus the entire organism.
The c-ring of a rotary ATPase is able to specifically bind either protons (H+) or sodium ions (Na+) as the coupling ion for the enzyme. Several structures are already available revealing the coordination network of both types of rotor rings. In each case ion binding includes a highly-conserved carboxylic acid residue (glutamate or aspartate), in addition to a more varying combination of amino acid residues, whereby Na+ coordination is structurally more demanding than H+ binding.
In the first part of my PhD thesis, I aimed to characterize the F1Fo ATP synthase rotor ring of the opportunistic pathogenic bacterium Fusobacterium nucleatum on a functional and structural level. F. nucleatum is an anaerobic bacterium which uses peptides and amino acids as a primary energy source. It is one of the most frequently occuring bacteria in human body infections and involved in human periodontal diseases.
The protein complex was heterologously expressed within a hybrid ATP synthase in Escherichia coli and purified without an affinity tag for further analysis. Two high resolution X-ray structures of the c-ring were solved at low (5.3) and high (8.7) pH to 2.2 and 2.64 Å, respectively. In both structures, the conserved glutamate is in an ion-locked conformation, revealing that the conformational state of the ion binding carboxylate is not depending on the pH of the crystallization condition, which is in good agreement with previous structural and biochemical studies of other c-rings.
A Na+ ion is present within the c-ring binding site and directly coordinated by four amino acid residues and a structural water molecule. Remarkably, the Na+ is bound by two glutamate residues instead of one as is the case in the I. tartaricus Na+ binding c-ring, of which the first high resolution X-ray structure of a c-ring has been solved in 2005. Thus, a new type of Na+ coordination in an ATP synthase rotor ring with a two-carboxylate ion binding motif is described here, which also occurs in other bacteria, including several pathogens. Na+ specificity of the investigated c-ring was further confirmed by a competitive biochemical labeling reaction performed with a fluorescent ATP synthase inhibitor molecule (N-cyclohexyl-N`-[4(dimethylamino)-α-naphtyl] carbodiimide, NCD-4).
We furthermore complemented our functional and structural data of the F. nucleatum c-ring by computational studies to explore the ion translocation mechanism of this enzyme in more details. We therefore analyzed the protonation state of the second, additional glutamate in the ion binding site. Molecular dynamics (MD) simulations and free-energy calculations indicated that this glutamate is constitutively protonated, in the ion-locked as well as in a simulated, more hydrated open-conformation of the ion binding glutamate as when it is travelling through the a/c-ring interface upon c-ring rotation.
G protein coupled receptors (GPCRs) constitute the largest family of cell-surface receptors in mammals and are key players in signal transduction. By responding to a plethora of extracellular stimuli ranging from photons to amines to fatty acids to peptides and proteins, these receptors trigger intracellular signalling cascades and regulate a variety of cellular responses. Approximately 800 genes in humans encode GPCRs which are classified according to sequence conservation into rhodopsin-like, glutamate, adhesion, frizzled/taste2 and secretin receptors. GPCRs share a seven transmembrane domain fold undergoing a conformational change upon ligand binding which is translated to the intracellular surface of the receptor thereby allowing a heterotrimeric G protein to couple. Heterotrimeric G proteins consist of a Ga, Gb and Gg subunit and dissociate into their Ga and Gbg entities upon activation by a GPCR. Subsequently, distinct signalling cascades are triggered by each G protein protomer.
Membrane proteins and GPCRs in particular, are highly important targets in drug design and development as currently approximately 60% of all marketed drugs target membrane proteins. Although these classes of proteins are of high therapeutic interest, our understanding of their mechanism of action and structure remains limited. The first structure of a human GPCR was determined in 2007 and required the development of protein engineering and innovative crystallisation techniques. Since then, approximately 130 GPCR structures of less than 40 individual receptors have been determined providing insights into the structural arrangement of the transmembrane helices, ligand binding pockets and G protein interactions. Combined with spectroscopic methods, these studies allowed a more detailed understanding of the molecular aspects of GPCR activation and signalling. Despite the tremendous advances in GPCR structural biology, certain aspects of GPCR function still remain poorly understood. Due to their size and inherent flexibility, the interaction of protein and peptide ligands with their receptors remains a challenging aspect in the structural characterisation of GPCRs. Moreover, structural information on subtype selectivity of peptide ligands continues to be scarce. To contribute functional and structural information on the molecular mechanisms of peptide interactions with GPCRs, this thesis focused on characterising receptors from the chemoattractant cluster using radioligand binding assays as well as NMR spectroscopy.
The chemoattractant cluster mainly groups the kinin, angiotensin, anaphylatoxin chemotactic complement and apelin receptors according to conserved residues in their ligand binding cavities. All receptors in this cluster bind to peptide ligands deriving from high molecular weight protein precursors upon proteolytic processing. Comparable to the conserved binding pocket of the chemoattractant receptors, the peptide ligands display a certain sequence conservation although they differ strongly in size. The largest ligands used in this thesis are the anaphylatoxins complement 3a and 5a, comprising 77 or 74 residues, respectively. Due to their size and complex fold involving three intramolecular disulphide bonds, solid phase synthesis is impossible, which prompted us to develop a modified cell-free expression system to produce these ligands in tritiated form for subsequent functional characterisation of the complement receptors. To demonstrate the versatility of the developed system, it was applied to another disulphidebond containing peptide ligand, the 21 amino acid endothelin-1. We describe a reliable and multifaceted tool to generate custom labelled peptide ligands for the structural and functional characterisation of GPCRs. The system allows the production of custom radioligands, peptides labelled for NMR studies or with fluorescent amino acids.
Apart from the modulation of GPCR activity by orthosteric ligands, GPCR signalling has long been described to be regulated by allosteric ligands including peptides, small molecules and ions. In this thesis, the influence of sodium ions on the activity state of the chemoattractant cluster receptors and in particular on the apelin, bradykinin 2 and angiotensin II type 1 receptors was examined. In recent high resolution crystal structures an allosteric sodium ion pocket beneath the orthosteric ligand binding cavity was identified and residues contributing to the coordination of sodium ions are conserved throughout the chemoattractant cluster receptors. This allosteric sodium ion coordinated within the transmembrane domain bundle has been described to negatively influence the affinity of agonists but not of antagonists. It was found that sodium ions have distinct influences on the affinity state as well as the available number of binding sites of the chemoattractant receptors. In case of the apelin and bradykinin 2 receptors, sodium ions drastically reduced the number of available binding sites whereas the affinity of peptide ligands to the bradykinin 2 receptors remained constant and the ligand binding affinities to the apelin receptor were completely abolished. In contrast, the angiotensin II type 1 receptor affinity state towards the endogenous peptide ligand angiotensin II is highly dependent on the presence of sodium ions, whereas binding of the synthetic peptide antagonist Sar1-Ile8-angiotensin II remained unaffected by the sodium ion concentration. As differential effects irrespective of the efficacy class but dependent on the amino acid composition of the applied ligands are observed, it can be concluded that electrostatic interactions between charged residues of the peptide ligands and amino acids on the extracellular surface of the receptors are influenced by sodium ions thereby adding another layer of complexity on GPCR signalling.
To elucidate the structure-function relationship of ligand selectivity between the kinin receptors, the structure of desArg10-kallidin (DAK) bound to the bradykinin 1 receptor was determined using solid state NMR (SSNMR) in the course of this thesis. The kinin peptides DAK and bradykinin bind with high affinity and high selectivity to either the bradykinin 1 or bradykinin 2 receptor, respectively. The binding pockets of the receptors are highly conserved and the two peptide ligands only differ in one amino acid at their N- and C-termini whereas the remaining eight amino acids are fully conserved. DAK adopts a U-shaped structure when bound to the bradykinin 1 receptor which resembles a horse shoe-like conformation. Using 2D TEDOR spectroscopy it could furthermore be demonstrated that positively charged residues at the N-terminal part of the peptide engage in ionic interactions with negatively charged amino acids on the extracellular surface of the bradykinin 1 receptor. In contrast, bradykinin displays a distinct b-turn at the C-terminus and an S-shaped conformation of the N-terminal segment when bound to the bradykinin 2 receptor. By using SSNMR to study the binding mode of DAK on the bradykinin 1 receptor we could determine that subtype selectivity between the kinin receptors is conferred by distinct conformational restraints within the peptide ligands and by the formation of specific ionic interaction between charged residues on the peptide and receptor, respectively.
In brief, this thesis contributes structural and functional data on the binding mechanisms and binding mode of different peptide-ligand GPCRs helping to understand subtype selectivity and allosteric modulation of the chemoattractant cluster receptors. In addition, a versatile cell-free expression system was developed that allows the custom synthesis of isotopically labelled peptides containing disulphide bonds for the functional characterisation of GPCRs.
Perioperative Blutungskomplikationen stellen, insbesondere für neurochirurgisch behandelte Patienten, eine schwerwiegende Komplikation dar und sind mit einer signifikant erhöhten Morbidität und Mortalität assoziiert [1]. Der perioperative Blutverlust ist einer der Hauptgründe für die Transfusion allogener Blutprodukte [2, 3]. Transfusionen von Erythrozytenkonzentraten (EK), Gefrorenem Frischplasma (GFP) und Thrombozytenkonzentraten (TK) sind dabei unabhängige Risikofaktoren für die Entwicklung thromboembolischer, pulmonaler, hämodynamischer oder septischer Komplikationen und selbst mit einer erhöhten perioperativen Morbidität und Mortalität assoziiert [3-10]. In der Literatur wird die Prävalenz von Blutungskomplikationen im Rahmen von intrakraniellen Eingriffen (Raten zwischen 0,8% bis 50%) sehr unterschiedlich beschrieben [1, 11-13]. Gleichwertige Voraussetzungen für die Bildung und Aufrechterhaltung eines stabilen Blutgerinnsels sind, neben einer effizienten primären Hämostase (Thrombozytenzahl und –funktion, Von-Willebrand-Faktor), auch die plasmatische Gerinnung (extrinsische und intrinsische Gerinnungsfaktoren) und der Schutz vor (Hyper)fibrinolyse [14].
The stable isotope geochemistry of pedogenic and lacustrine carbonate and fossil herbivore tooth enamel is a powerful tool to study the evolution of terrestrial paleoenvironments. This thesis aims to reconstruct Neogene ecosystems in the Karonga Basin in the southern part the East African Rift (EAR) and the Central Anatolian Plateau (CAP).
Karonga Basin: Understanding the development of East African savanna biomes is crucial for reconstructing the evolution, migration and dietary behaviors of early hominins. These rift ecosystems range from closed woodland to open grassland savanna and vary widely in fraction of woody cover. Here I present Plio-Pleistocene carbon (δ13C) and oxygen (δ18O) records from pedogenic carbonate and fossil herbivore tooth enamel collected from the Malawi Rift. This is the first southern hemisphere long-term record in the East African Rift.
The studied ca. 4.3 Ma to 0.6 Ma deposits of the Chiwondo Beds (Karonga Basin) are home to two hominin fossil finds, a maxillary fragment of Paranthropus boisei and a mandible of Homo rudolfensis, both dated to ca. 2.4 Ma. The study site is situated between the well-known hominin-bearing localities of eastern and southern Africa and hence fills an important geographical gap for early hominin research.
The δ13C values of pedogenic carbonate (n = 321) and of enamel from 14 different large-bodied herbivorous mammal taxa (suid, equid, bovid, elephant an hippopotamus; n = 122) permit assessment of the evolutionary history of C3 and C4 biomass, which is closely linked to climate patterns. The reconstruction of C4-grassland development offers insights into the retreat of tree cover and the distribution and seasonality of precipitation. δ18O values reflect hydrological patterns, and we supplement the data of fossil proxy material with analyses of modern meteoric water to evaluate influences of seasonality and evaporation.
Consistent δ13C values around -9‰ and δ18O values of ca. 24‰ of the pedogenic carbonate from 14 sections spanning the last 4.3 Ma indicate a relatively mesic and persistent climate with C3-dominated woodland savannas in the Karonga Basin. The data from tooth enamel of suid, elephant, and hippo taxa complement these findings with constantly low δ13C and δ18O values, demonstrating intake of mainly C3 biomass and water from sources which are only very limited affected by evaporation.
In contrast, Karonga Basin equid and bovid taxa reflect much more complex dietary patterns, ranging from mixed C3/C4-feeders to strongly C4-influenced diets. Intra-tooth δ18O values also show large variations, indicating water intake from resources that differ in their isotopic composition and therefore experienced different hydrological settings.
Overall, the stable isotope data reflect a mostly C3-dominated mesic paleoecosystem, which comprises patches of more open C4-grasslands within the range of migrating mammals. Supplementary analyses of δ18O values of modern meteoric water (precipitation, lake, river and groundwater; n = 111) show a similar magnitude to the fossil herbivore oxygen isotopic values. Expected 18O values of the drinking water calculated from herbivore enamel of animals with a mixed- or C3-diet indicate water intake from barely evaporated sources. Specialized feeders with a high C4-consumtion, however, reflect s|||||||||||||||||||||values of 16O-depleted reservoirs, similar in δ18O |||||||||||| to modern evaporated lakes.
The absence of long-term trends towards more positive δ13C and δ18O values in the Karonga Basin contrasts the increasing role of C4-grasslands since ca. 2.5 Ma in the Eastern Rift. Our data hence point to regional differences in climate and vegetation dynamics during the Plio-Pleistocene. Therefore, regions that are home to early hominins such as H. rudolfensis and P. boisei may have had a different environmental history when compared to the Eastern Rift. This suggests that hominin adaptation is not necessarily directly linked to the emergence of open landscapes.
Central Anatolian Plateau: The development of relief and climate patterns in the Central Anatolian Plateau is long-standing debate in modern paleoecological studies. This thesis presents long-term δ13C and δ18O records on five lacustrine successions, which are widely distributed across the CAP. Also, new 40Ar/39Ar geochronological data from volcanic ashes are established in addition to existing biostratigraphic and paleomagnetic data.
Field relationships combined with stable isotope data of 230 lacustrine carbonates indicate a Late Oligocene environment that was characterized by large, temporally open freshwater lakes in a relatively humid subtropic climate. In the middle Aquitanian, an increase in lake δ18O values reflects more arid settings and an overall increasing dominance of closed saline lakes. This time was probably characterized by frequent climatic fluctuations, recording the influence of seasonality, topography and the waxing and waning of aridity.
Die 5-Lipoxygenase (5-LOX) stellt den Startpunkt des Leukotrienstoffwechsels dar, da sie Arachidonsäure (AA) über die 5(S)-Hydroperoxy-6-trans-8,11,14-cis-eicosatetraensäure (5-HpETE) in Leukotrien A4 (LTA4) umwandelt. 5-HpETE kann zum korrespondierenden Alkohol 5(S)-Hydroxy-6-trans-8,11,14-cis-eicosatetraensäure (5-HETE) reduziert werden. LTA4 dient als Zwischenprodukt für die Synthese von LTB4 und den Cysteinyl-gebundenden LTs LTC4, LTD4 und LTE4. LTs nehmen eine wichtige Funktion in der Immunabwehr ein, sind jedoch auch an einer Vielzahl von Krankheitsgeschehen wie z. B. Asthma bronchiale, Atherosklerose und einiger Tumorarten beteiligt. Die 5-LOX teilt sich in zwei Domänen auf: der reglatorischen, N-terminalen Domäne und der katalytischen, C-terminalen Domäne. Ihre Aktivität unterliegt einer komplexen allosterischen Regulation und kinetischen Besonderheiten wie einer Substratinhibition. In vielen Fällen ist die regulatorische PLAT-(Polycystin-1, Lipoxygenase, alpha-Toxin)-Domäne involviert. Sie ist essentiell an der Bindung von Calcium, Membranen und weiterer Faktoren wie dem Coactosin-like protein (CLP) und Dicer beteiligt. Auch eine zweite Bindungsstelle für das Substrat oder einen seiner Metaboliten wird dort vermutet. Letztlich bleibt jedoch die Regulation der 5-LOX-Aktivität durch die PLAT-Domäne unzureichend geklärt. Diese Tatsache und die fortwährende Suche nach neuen Ansatzpunkten für die 5-LOX-Inhibition bilden den Hintergrund, vor dem diese Arbeit angefertigt wurde.
Das Ziel lag in der Entwicklung einer stabilen, isolierten PLAT-Domäne und deren Charakterisierung. Es stellte sich jedoch heraus, dass sich die isolierte Domäne durch eine hohe thermische Instabilität und starke Aggregationsneigung auszeichnet. Mittels Mutationsstudien auf Basis der 5-LOX AS 1-115, verbunden mit Gelfiltrationsläufen zur Analyse der Proteinaggregation, wurde schließlich ein Konstrukt entwickelt, das in Konzentrationen < 0,5 mg/ml als Monomer vorlag: die sogenannte PLAT1-115 W75G. Ein Austausch des W75 in Glycin erhöhte ebenfalls die thermische Stabilität, so dass Versuche bei 20°C durchgeführt werden konnten. Zunächst wurden jedoch die grundlegenden Eigenschaften der Mutante untersucht. Dies umfasste die Beantwortung der Frage, ob auch die PLAT1-115 W75G Calcium bindet, sowie die Aufnahme eines Circulardichroismus-(CD)-Spektrums. Der erste Aspekt konnte mit mehreren Methoden bestätigt werden. Eine Calciumzugabe zum Laufpuffer 20 mM MOPS, 50 mM KCl pH 7,4 erhöhte konzentrationsabhängig das Elutionsvolumen der PLAT1-115 W75G auf der analytischen Gelfiltrationssäule – vermutlich durch den bekannten Einfluss von Calcium auf die Hydrophobizität der PLAT-Domäne. Zusätzlich wurde die Interaktion durch differential scanning fluorimetry (DSF) und Oberflächen-Plasmonen-Resonanz-Spektroskopie (SPR) nachgewiesen. Allerdings gelang aus verschiedenen Gründen keine Quantifizierung der Bindungsaffinität. Das CD-Spektrum bestätigte die Struktur der PLAT-Domäne als sogenanntes all-beta_protein und ermöglichte die Einordnung der PLAT1-115 W75G in die Gruppe der betaII-Proteine.
Ein weiterer Fokus dieser Arbeit lag auf der vermuteten allosterischen Fettsäurebindungsstelle in der PLAT-Domäne. Es wurde versucht, die Interaktion mittels SPR nachzuweisen. Zur Vorbereitung wurde im 5-LOX-Aktivitätstest und im DSF an der isolierten Domäne ein Detergens bestimmt, das einen möglichst geringen Einfluss auf das Protein ausübt. Dabei zeigte Octyl-beta-D-glucopyranosid (beta-OG) das vorteilhafteste Profil. Auf dieser Basis wurde die kritische Mizellbildungs-Konzentration (CMC) der AA und einiger HETEs in beta-OG-haltigen Puffern bestimmt. Die SPR-Studien ergaben jedoch keine reproduzierbaren Ergebnisse. In einem weiteren Schritt wurden die Substrathemmung des Gesamtproteins 5-LOX und der Einfluss von Calcium charakterisiert. Sowohl in Gegenwart von ~ 1 mM freiem Calcium als auch von 1 mM EDTA lag mit 20 µM AA die höchste Produktbildung nach 10-minütiger Reaktion vor. Das Detergens Tween20 (T20) hob in einer Konzentration unter seiner CMC (0,001 % m/V) in Anwesenheit von Calcium die Inhibition auf. Ohne Calcium zeigte sich auch in Gegenwart von T20 die bekannte Substratinhibition der 5-LOX einschließlich ihrer Maximalaktivität bei 20 µM AA. Diese Ergebnisse deuten darauf hin, dass Calcium eine Bindung der 5-LOX an eventuell vorhandene, negativ geladene Vesikel aus AA und Detergens vermitteln und dadurch die Substratinhibition aufheben kann. In Fällen, in denen die Substratinhibition vor dem Erreichen der AA-CMC auftritt, hat Calcium folglich keinen Einfluss.
Zuletzt wurde die Interaktion der PLAT1–115 W75G mit CLP und einem C-terminalen Fragment von Dicer untersucht. Im Crosslinking ließ sich nicht auf eine Interaktion der isolierten PLAT-Domäne mit CLP schließen. Dagegen ergaben Diamid-Crosslinking-Studien, dass die isolierte PLAT-Domäne in der Lage ist, das Dicer-Fragment zu binden. Dieses Ergebnis wurde im SPR bestätigt.
Sowohl bezüglich der Modellierung der Sprachkompetenz bei mehrsprachigen Kindern als auch hinsichtlich der Bestimmung der hierfür notwendigen Indikatoren herrscht nach wie vor Forschungsbedarf. Die entsprechenden Erkenntnisse sind aber für eine valide Sprachstandsdiagnostik, auf Basis welcher auch eine Sprachförderung stattfinden kann, unerlässlich. In der vorliegenden Dissertation werden daher mit Hilfe von Verteilungsstatistiken, konfirmatorischen Faktoranalysen und Strukturgleichungsmodellen zunächst ausgewählte Sprachstandsindikatoren sowie das Konstrukt Sprachkompetenz von Kindern mit Deutsch als Zweitsprache zu Beginn der 1. Klasse modelliert und im Anschluss ihr Einfluss auf die Orthographiekompetenz dieser Schülerinnen und Schüler am Ende der 2. Klasse untersucht. Zusätzlich wird das phonologische Arbeitsgedächtnis als ein weiterer Prädiktor in das Modell eingebunden. Es wird der Fragestellung nachgegangen, ob die für die Modellierung der Sprachkompetenz verwendeten Indikatoren, wie bspw. Bildbenennung, Kasus, Syntaxerwerbsstufen und die mittlere Äußerungslänge (MLU) für die Gruppe der mehrsprachigen Kinder in der Schuleingangsphase geeignet bzw. valide sind. Dabei werden die jeweiligen Streuungen, die Korrelationen untereinander (konvergente Validität) sowie die Leistung der Indikatoren für die jeweiligen sprachlichen Kompetenzbereiche anhand der Höhe der Faktorladungen betrachtet. Ebenfalls wird die prognostische Validität der Indikatoren hinsichtlich der Rechtschreibkompetenz beleuchtet. Des Weiteren wird geprüft, ob sich die Sprachkompetenz bei DaZ-Kindern in eine semantische, eine morphologische und eine syntaktische Fähigkeit unterteilen lässt und ob die Sprachkompetenz als Faktor zweiter Ordnung modelliert werden kann. Weitere Fragestellungen betreffen die Modellierung der Rechtschreibkompetenz, also die Anzahl und Art der latenten Variablen, die dieses Konstrukt abbilden, sowie den Einfluss des phonologischen Arbeitsgedächtnisses und der einzelnen sprachlichen auf die orthographischen Teilkompetenzen.
Ein Finite-Volumen-Modell des differentiell geheizten rotierenden Annulus wird verwendet, um die spontane Schwerewellenabstrahlung durch die großskalige, von baroklinen Wellen beherrschte Strömung zu untersuchen. Bei diesem Vorgang bilden barokline Wellen und der durch sie abgelenkte und verzerrte Strahlstrom, die sich näherungsweise im hydrostatischen und geostrophischen Gleichgewicht befinden, durch ihre Dynamik Ungleichgewichte aus, die sich als Schwerewellen ausbreiten. Neben der Anregung von Schwerewellen durch Prozesse wie Gebirgsüberströmung, Konvektion und Frontogenese, bildet dieser Vorgang vermutlich eine weitere wichtige Quelle von Schwerewellen in der Atmosphäre. Anders als für orographisch und konvektiv angeregte Schwerewellen gibt es für die spontane Schwerewellenabstrahlung bislang keine befriedigende Parametrisierung in Wettervorhersage- und Klimamodellen, die diesen Prozess nicht auflösen können. Die Durchführung von Messungen zur spontanen Schwerewellenabstrahlung in der Atmosphäre ist üblicherweise sehr aufwendig, sodass die Untersuchung dieses Vorganges in einem wiederholbaren und steuerbaren Laborexperiment reizvoll erscheint. Ob dafür möglicherweise das Experiment des differentiell geheizten rotierenden Annulus infrage kommt, untersuchen wir mit einem eigens dafür entwickelten numerischen Modell, dessen Tauglichkeit wir zunächst im Rahmen einer Validierung durch den Vergleich mit Labormessungen überprüfen. Damit die Ergebnisse zur Schwerewellendynamik im Annulus auf die Atmosphäre übertragbar sind, verwenden wir eine neue, atmosphärenähnliche Annuluskonfiguration. Im Gegensatz zu den klassischen Annuluskonfigurationen ist in der neuen Konfiguration die Brunt-Väisälä-Frequenz größer als der Coriolis-Parameter, sodass die Schwerewellen ein ähnliches Ausbreitungsverhalten zeigen sollten wie in der Atmosphäre. Deutliche Hinweise auf eine Schwerewellenaktivität in der atmosphärenähnlichen Konfiguration geben die horizontale Geschwindigkeitsdivergenz und eine Normalmodenzerlegung der kleinräumigen Strukturen der simulierten Strömung. Um der Herkunft der beobachteten Schwerewellen auf den Grund zu gehen, zerlegen wir die Strömung in den schwerewellenfreien quasigeostrophischen Anteil und den schwerewellenenthaltenden ageostrophischen Anteil. Bereiche innerhalb der baroklinen Welle, in denen ein erhöhter spontaner Antrieb des ageostrophischen Anteils durch die quasigeostrophische Strömung beobachtet wird, fallen mit Bereichen erhöhter Schwerewellenaktivität zusammen. Dies deutet darauf hin, dass die spontane Schwerewellenabstrahlung auch im Annulus zum Schwerewellenfeld beiträgt, sodass dieses Experiment als Labormodell dieser Schwerewellenquelle für deren weitere Erforschung geeignet erscheint.
Small molecule inhibitors sensitize neuroblastoma cells for chemotherapeutic drug-induced apoptosis

Neuroblastoma (NB) is one of the most common solid extracranial pediatric tumors, deriving from undifferentiated cells of the peripheral nervous system. It accounts for approximately 10% of all childhood cancers. High stage tumors usually show poor prognosis despite aggressive treatment such as radiotherapy or chemotherapy. Therefore, it is of utmost importance to find novel treatment strategies in order to improve existing chemotherapy protocols. Combination treatment offers advantages, as chemotherapeutic drugs can be applied in low and subtoxic doses, reducing possible side-effects. Here, we report in a two-part study that small molecule inhibitors (SMI), namely BI 2536, a PLK1 inhibitor and BV6, a SMAC mimetic (SM), sensitize neuroblastoma cells for chemotherapeutic drug-induced cell death. By using i) BI 2536 in combination with vinca alkaloids and ii) BV6 in combination with either doxorubicin or vinca alkaloids, we show that cell death is synergistically enhanced compared to monotherapy. Furthermore, combination treatment significantly reduces survival of NB cells in long-term assays, compared to single treatment. We identify that vinca alkaloid/SMI combinations induce mitotic arrest, as shown by phosphorylation of histone H3, which results in the induction of intrinsic apoptosis and inhibition of CDK1 by RO-3306 could abolish these findings. Mechanistically, upon vinca alkaloid/SMI-induced mitotic arrest, anti-apoptotic BCL-2 proteins such as MCL-1, BCL-2 or BCL-XL are degraded or inactivated by phosphorylation, which induces the activation of the proapoptotic BCL-2 family proteins BAX and BAK. The importance of the mitochondrial apoptosis pathway in vinca alkaloid/SMI-induced cell death was further highlighted by the fact that ectopic expression of BCL-2 inhibits vinca alkaloid/SMI-induced DNA fragmentation and BAK- and caspase-activation. In contrast to the vinca alkaloid/SMI cotreatment, DOX/SMI (DOX/BV6)-induced apoptosis only partially involves the mitochondrial pathway. Instead, we clarify that RIP1 is required for DOX/BV6-induced apoptosis, as pharmacological and genetic inhibition of RIP1 rescues from apoptosis induction. Although it has been shown in previous studies that SM-treatment (e.g. BV6) can induce the NF-κB pathway and auto-/paracrine TNFα production through cIAP1/2 depletion, DOX/BV6-induced apoptosis is completely independent of NF-κB activation in our setting, despite fast cIAP1 depletion. This conclusion is based on the fact that inhibition of the NF-κB pathway by exogenously expressed dominant-negative IκBα as well as application of a TNFα blocking antibody does not reduce DOX/BV6-induced cell death. In summary, we unravel two new promising treatment strategies for neuroblastoma patients by using a combination treatment of two different small molecule inhibitors, combined with well-characterized chemotherapeutic agents. Furthermore we give detailed insights into cell death pathways induced by these combination treatments, in which mitochondria and RIP1 have a differential role in chemotherapeutic drug-induced apoptosis.
Resistance in glucocorticoid-induced apoptosis is associated with poor prognosis for long term survival in childhood acute lymphoblastic leukemia (ALL). As Smac mimetics have been shown to reactivate apoptosis by antagonizing Inhibitor of Apoptosis (IAP) proteins, we investigate the potential of the Smac mimetic BV6 to overcome glucocorticoid-resistance in ALL. This study shows that BV6 synergistically cooperates with glucocorticoids to trigger apoptosis and to suppress clonogenic growth of pediatric ALL cells. Of note, the BV6/glucocorticoid combination treatment also induces cell death in cells having defects in the apoptotic signaling cascade by inducing a switch from apoptotic to necroptotic cell death. The clinical relevance of our novel combination treatment is underscored by parallel experiments in primary pediatric ALL samples, in which glucocorticoids and BV6 act together to induce cell death in a synergistic manner. Importantly, the addition of BV6 enhances the anti-leukemic effects of glucocorticoids in an in vivo mouse model of pediatric ALL without causing substantial side effects, highlighting the potency of a BV6/glucocorticoid combination treatment. In contrast, BV6 does not increase cytotoxicity of glucocorticoids against several non-malignant cell types of the lympho-hematopoietic system. Furthermore, we have identified the novel underlying mechanism of BV6/glucocorticoid-induced apoptosis by showing that BV6 and glucocorticoids synergistically act together to promote assembly of the ripoptosome, a RIP1/FADD/caspase-8-containing cell death complex. Ripoptosome assembly is critically required for BV6/Dexamethasone-induced cell death, since genetic silencing of its members, i.e. RIP1, reduces ROS production, caspase activation and most importantly cell death induction. BV6/glucocorticoid combination treatment promotes ripoptosome assembly by inhibition of both of its negative regulators, IAP proteins and cFLIP. Thus, we identify that BV6 and glucocorticoids cooperate together to reduce cIAP1, cIAP2 and XIAP protein levels and cFLIP expression. Ripoptosome formation occurs independently of autocrine/paracrine loops of death receptor ligands, since blocking antibodies for TNFα, TRAIL or CD95L or genetic silencing of their corresponding receptors fail to rescue BV6/glucocorticoid-induced cell death. In summary, this study shows that the Smac mimetic BV6 sensitizes for glucocorticoid-induced apoptosis by promoting ripoptosome assembly with important implications for the treatment of childhood ALL.
The PANDA experiment at FAIR will study fundamental questions of strong interaction with high precision. Effcient particle identification for a wide momentum range and the full solid angle is required for successful reconstruction of the benchmark channels of the broad PANDA physics program. For this purpose a compact ring imaging Cherenkov detector is being developed for the barrel region of the PANDA detector. The concept and the baseline design of the PANDA Barrel DIRC were inspired by the BABAR DIRC and improved with important modifications, like fast photon timing, a compact expansion volume, and focusing optics. The required detector resolution was defined based on the PANDA PID specifications using the phase space distributions of the final state kaons produced in selected benchmark channels. To optimize the PANDA Barrel DIRC design in terms of performance and cost the baseline detector geometry and a number of design options were implemented in the simulation. The key options include the radiator dimensions, two types of expansion volume shapes, and a variety of focusing systems. The performance of the detector designs was quantified in terms of single photon Cherenkov angle resolution and photon yield. It was found that the number of radiators can be reduced by about 40% without loss in performance. A compound spherical lens without air gap was found to be a promising focusing system. An optimized Barrel DIRC design meeting the PID requirements includes three radiator bars per at section, the compound lens without air gap, a compact prism-shaped EV, and a total of 192 Microchannel-Plate PMTs as photosensors. The number of electronic channels can be halved without loss in performance by combining two neighbouring pixels. For such a detector design the total cost will be significantly reduced compared to the baseline version while still meeting or exceeding the PANDA PID performance goals.
International society consists of states and the rules and institutions they share. Although international society has become a mundane feature of the world and the principal research focus of International Relations, it has become meaningless. More specifically, the technical rules that determine what states are and how they relate to other features of the world are units of semantic meaning, but their rampant, unprincipled proliferation has corroded their capacity to contain existential meaning. This deterioration is to be deplored because it alienates subjects from each other, it is totalising and excludes alternatives, and it is theoretically irreversible. To connect the two kinds of meaning, the first step is to reconceptualise international society as consisting strictly of constitutive rules whose meaning depends on the context they jointly compose, which implies that these rules can in turn be represented as signs in a semiotic structure. In order to evaluate the capacity of the signs to contain existential meaning, the next step is to adapt Baudrillard’s hierarchical typology of semiotic systems, ranging from the most meaningful systems based on symbolic exchange value to the vapid terminus of hyperreality based on sign value, in which semantic meaning is without value and existential meaning is impossible. The narrative traces the history of the signs of international law from the premodern period, when Christendom was understood as an approximation of the divine kingdom and a vehicle for salvation, to the present postmodern period, in which hundreds of articles of international maritime law make the decision to go to war over isolated rocks intelligible – even rational – and international trade law catalogues potato products to six digits. Three cases in particular exemplify this devolution in international law: the laws determining the territorial sea, the most-favoured national principle of international trade law, and nationality as a normative basis for statehood.
Die Dissertation ist in den Bereichen der semiklassischen Quantengravitation und der pseudokomplexen Allgemeinen Relativitätstheorie (pk-ART) anzusiedeln. Dabei wird unter semiklassischer Quantengravitation die Untersuchung quantenmechanischer Phänomene in einem durch eine klassische Gravitationstheorie gegebenen gravitativen Hintergrundfeld verstanden und bei der pk-ART handelt es sich um eine Alternative zu der aktuell anerkannten klassischen Gravitationstheorie, der Allgemeinen Relativitätstheorie (ART), die die reellen Raumzeitkoordinaten der ART pseudokomplex erweitert. Dies führt zusammen mit einer Veränderung des Variationsprinzips in führender Ordnung auf eine Korrektur der Einstein- Gleichung der ART mit einem zusätzlichen Quellterm (Energie-Impuls-Tensor), dessen exakte Form jedoch bisher nicht bekannt ist.
Die Beschreibung der Gravitation als Hintergrundfeld ergibt sich zwangsläufig daraus, dass auf Basis der ART bisher keine quantisierte Beschreibung für sie gefunden werden konnte. Jedoch wird erhofft, dass die Untersuchung semiklassischer Phänomene Hinweise auf die korrekte Theorie der Quantengravitation gibt. Zudem motiviert der Mangel einer quantisierten Gravitationstheorie die Verwendung alternativer Theorien, da sich dadurch die Frage stellt, ob die ART die korrekte Beschreibung klassischer Felder ist.
Das Ziel der vorliegenden Dissertation war die grundlegenden Unterschiede zwischen der ART und der pk-ART für gebundene sphärisch symmetrische Zustände der Klein-Gordon- und der Dirac-Gleichung zu identifizieren und ein qualitatives Modell der Vakuumfluktuationen in sphärisch symmetrischen Materieverteilungen zu bestimmen, wobei der Zusammenhang der pk-ART mit den Vakuumfluktuationen in der Annahme besteht, dass ein Zusammenhang zwischen ihnen und dem zusätzlichen Quellterm der pk-ART existiert. Dafür wurden die gebundenen Zustände der Klein-Gordon- und der Dirac-Gleichung für drei verschiedene Metrikmodelle (zwei ART-Modelle und ein pk-ART-Modell) mit konstanter Dichte systematisch numerisch berechnet, einige repräsentative Grafiken erstellt, anhand derer die grundlegenden Unterschiede der Ergebnisse der ART-Modelle und des pk-ART-Modells erörtert wurden, und die ART Ergebnisse der Dirac-Gleichung soweit wie möglich mit Ergebnissen der Literatur verglichen. Insbesondere wurde dabei festgestellt, dass die Energieeigenwerte in der pk-ART im Gegensatz zu denen in der ART in Abhängigkeit der Ausdehnung des Zentralobjekts ein Minimum aufweisen. Zudem wurden die Energieeigenwerte der Klein-Gordon-Gleichung teilweise sowohl über das Eigenwertproblem einer Matrix als auch über ein Anfangswertproblem berechnet und es wurde festgestellt, dass die Beschreibung als Eigenwertproblem deutlich uneffektiver ist, wenn dafür die Basis des dreidimensionalen harmonischen Oszillators genutzt wird. Für die Entwicklung des qualitativen Vakuumfluktuationsmodells wurden zwei Näherungen für den Erwartungswert des Energie-Impuls-Tensors in führender Ordnung für die Schwarzschildmetrik (ART) verglichen und die Verwendung eines qualitativen Modells durch die dabei auftretende Diskrepanz gerechtfertigt. Danach wurden die Vakuumfluktuationen für Metriken konstanter Materiedichte mit Hilfe einer der Näherungen in führender Ordnung berechnet und ein Modell gesucht, das den gleichen qualitativen Verlauf aufweist. Im Anschluss wurde dieses Modell noch für einfache Metriken mit variabler Materiedichte verifiziert.
Die Dissertation leistet mit der Analyse der gebundenen Zustände einen Beitrag in der Identifikation der Unterschiede zwischen der pk-ART und der ART und führt somit auf weitere mögliche Messgrößen, die der Unterscheidung der beiden Theorien dienen könnten. Weiterhin ermöglicht das abgeleitete Modell eine Verfeinerung der schon publizierten Ergebnisse über Neutronensterne und die für die Erstellung nötigen Vorarbeiten leisten einen Beitrag zur Identifikation des
pk-ART Quellterms.
Die vorliegende Arbeit verfolgt den Anspruch, die von Paul Natorp (1907) gestellte Frage, was die Gemeinschaft für die Erziehung und umgekehrt die Erziehung für die Gemeinschaft bedeute, empirisch auszuloten. Im Mittelpunkt des Forschungsinteresses steht dabei das Verhältnis von Schule und Gemeinschaft in der ‚postnationalen Konstellation‘ (Habermas 1998), dem sich die Arbeit über die Untersuchung von Unterrichtskommunikation zu den Themen ‚Nationalsozialismus/Holocaust‘ und ‚Multikulturalismus/Rassismus‘ anzunähern versucht.
Zur Vorbereitung der empirischen Studie wird im Rahmen einer Semantikstudie zunächst der pädagogische Diskurs zum Thema Gemeinschaft von den ersten Vorläufern gemeinschaftspä-dagogischen Denkens bis hin zu aktuellen konzeptionellen Entwürfen nachgezeichnet. Funktion und Bedeutung der Gemeinschaftsfigur als Bezugskategorie von Erziehungsreflexion in der Moderne werden herausgearbeitet. Weiterhin werden die rekonstruierten gemeinschaftspädagogischen Konzepte in Hinblick auf ihr Spannungspotential zu Leitprinzipien demokratischer Erziehung beleuchtet.
Mithilfe sequenzanalytischer Interpretationen wird anschließend der Frage nachgegangen, welche Rolle Gemeinschaftsbezügen als Fluchtpunkt pädagogischer Einwirkungsbemühungen im Unterricht zukommt. Gleichzeitig wird gefragt, ob und in welcher Weise Unterricht Gemeinschaft als Ermöglichungsform zur Umsetzung seiner pädagogischen Absichten in Anspruch nimmt. Nicht zuletzt richtet die Analyse das Augenmerk auf den Umgang mit den potentiellen Herausforderungen, die unter Migrationsbedingungen mit dem Rekurrieren auf Gemeinschaft am Lernort (Geschichts-)Unterricht verbunden sind.
Die Interpretationen zeigen, wie Unterricht Gemeinschaft in Anspruch nimmt, um die Bedingungen seines Prozessierens zu sichern. Weiterhin decken sie zwei kontrastierende Typen des Rekurrierens auf Gemeinschaft als Fluchtpunkt pädagogischer Kommunikation zum Thema NS auf. Als Lösungsoption im Umgang mit den migrationsbedingten Herausforderungen von Geschichtsunterricht zum Thema NS deutet sich indes die Tendenz an, verstärkt auf eine universalistische Erinnerungs- und Verantwortungskultur Bezug zu nehmen.
Das Gefahrenpotential, das dem Gemeinschaftsgedanken in der aktuellen erziehungswissenschaftlichen Diskussion zugeschrieben wird, erweist sich in den betrachteten Unterrichtsse-quenzen als beschränkt. Die Beobachtungen legen vielmehr die These einer eingehegten Form von Gemeinschaftserziehung in der gegenwärtigen Unterrichtspraxis nahe. Darin erfährt das Risiko, das gemeinschaftspädagogischen Ansätzen ihrer Kritik nach eingeschrieben ist, über normative Selbstverpflichtungen der pädagogischen Praxis eine Eindämmung.
Acute myeloid leukemia is a hematopoietic stem cell disorder and a type of acute leukemia which is characterized by clonal proliferation of myeloid precursors with a reduced capacity to differentiate into more mature cellular elements. Clinically AML is characterized by a high degree of heterogeneity with respect to chromosome abnormalities, gene mutations, and changes in expression of multiple genes and microRNAs. Cytogenetic abnormalities can be detected in approximately 50% to 60% of newly diagnosed AML patients. Majority of AML cases are associated with chromosomal aberrations, more specifically translocations that often result in gene arrangements and expression of aberrant fusion proteins. This study was carried out with two fusion proteins: PML/RARα and DEK/CAN which results from the translocations t(15;17) and t (6,9) respectively. PML/RARα is the most common translocation (97%) and the main driver in Acute Promyelocytic Leukemia (APL), a wellcharacterized and well treatable subtype of AML. In contrast, DEK/CAN occurs in 1-5% of AML, associated with poor prognosis and defines a high risk group in AML. The expression of PML/RARα results in a fusion protein that acts as a transcriptional repressor by interfering with gene expression programs involved in differentiation, apoptosis, and selfrenewal. Current therapy focused on the targeting of PML/RARα fusion protien. Success has been achieved by using either ATRA, anthracyclines and Arsenic trioxide or their combinations. These agents induce differentiation in PML/RARα positive AML and hence called differentiation therapy. In comparison with ATRA, ATO and anthracyclines are poor cellular differentiation agents. Despite early promise, several studies have reported that differentiation therapy is unable to target/eradicate leukemic stem cells or eradicate the disease. Therefore current therapeutic focus is to eliminate leukemic stem cells and achieve complete molecular remission not only in APL but also in acute lymphoblastic leukemia and chronic myeloid leukemia as well. Key enzymes of the eicosanoid pathways in the arachidonic acid metabolism, such as COX1/2 as well as the 5-LO have been shown to be good targets for leukemic stem cell therapy approach in AML by interfering with the Wntsignaling which is known to be indispensable for the pathogenesis of AML.
