Chemokines play a key role in the cellular infiltration of inflamed tissue. They are released by a wide variety of cell types during the initial phase of host response to injury, allergens, antigens, or invading microorganisms, and selectively attract leukocytes to inflammatory foci, inducing both migration and activation. Monocyte chemoattractant protein-1 (MCP-1), a member of the CC chemokine superfamily, functions in attracting monocytes, T lymphocytes, and basophils to sites of inflammation. MCP-1 is produced by monocytes, fibroblasts, vascular endothelial cells and smooth muscle cells in response to various stimuli such as tumour necrosis factor-a (TNF-a), interferon-g (IFN-g), and interleukin-1b (IL-1b). It also plays an important role in the pathogenesis of chronic inflammation, and overexpression of MCP-1 has been implicated in diseases including glomerulonephritis and rheumatoid arthritis. Oligonucleotide-directed triple helix formation offers a means to target specific sequences in DNA and interfere with gene expression at the transcriptional level. Triple helix-forming oligonucleotides (TFOs) bind to homopurine/homopyrimidine sequences, forming a stable, sequence-specific complex with the duplex DNA. Purine-rich sequences are frequent in gene regulatory regions and TFOs directed to promoter sequences have been shown to prevent binding of transcription factors and inhibit transcription initiation and elongation. Exogenous TFOs that bind homopurine/ homopyrimidine DNA sequences and form triple-helices can be rationally designed, while the intracellular delivery of single-stranded RNA TFOs has not been studied in detail before. In this study, expression vectors were constructed which directed transcription of either a 19 nt triplex-forming pyrimidine CU-TFO sequence targeting the human MCP-1 or two different 19 nt GU- or CA-control sequences, respectively, together with the vector encoded hygromycin resistance mRNA as one fusion transcript. HEK 293 cells were stable transfected with these vectors and several TFO and control cell lines were generated. Functional relevant triplex formation of a TFO with a corresponding 19 bp GC-rich AP-1/SP-1 site of the human MCP-1 promoter was shown. Binding of synthetic 19 nt CUTFO to the MCP-1 promoter duplex was verified by triplex blotting at pH 6.7. Underlining binding specificity, control sequences, including the GU- and CA-sequence, a TFO containing one single mismatch and a MCP-1 promoter duplex containing two mismatches, did not participate in triplex formation. Establishing a magnetic capture technique with streptavidin microbeads it was verified that at pH 7.0 the 19 nt TFO embedded in a 1.1 kb fusion transcript binds to a plasmid encoded MCP-1 promoter target duplex three times stronger than the controls. Finally, cell culture experiments revealed 76 ± 10.2% inhibition of MCP-1 protein secretion in TNF-a stimulated CU-TFO harboring cell lines and up to 88% after TNF-a and IFN-g costimulation in comparison to controls. Expression of interleukin-8 (IL-8) as one TNF-a inducible control gene was not affected by CU-TFO, demonstrating both highly specific and effective chemokine gene repression. Furthermore, another chemokine target, regulated upon activation normal T cell expressed and secreted (RANTES), which plays an essential role in inflammation by recruiting T lymphocytes, macrophages and eosinophils to inflammatory sites, was analysed using the triplex approach. A 28 nt TFO was designed targeting the murine RANTES gene promoter, and gel mobility shift assays demonstrated that the phosphodiester TFO formed a sequencespecific triplex with the double-stranded target DNA with a Kd of 2.5 x 10-7 M. It was analysed whether RANTES expression could be inhibited at the transcriptional level testing the TFO in two different cell lines, T helper-1 lymphocytes and brain microvascular endothelial cells (bend3 cells). Although there was a sequence-specific binding of the TFO detectable in the gel shift assays, there was no inhibitory effect of the exogenously added and phosphorothioate stabilised TFO on endogenous RANTES gene expression visible. Additionally, the small interfering RNA (siRNA) approach was tested as another strategy to inhibit expression of the pro-inflammatory chemokines MCP-1 and RANTES. Two different methods were pursuit, describing transient transfection with vector derived and synthetic siRNA. The vector pSUPER containing the siRNA coding sequence was used to suppress endogenous MCP-1 in HEK 293 cells. An empty vector without RNA sequence served as a control. Inhibition due to the siRNA was measured in stimulated and unstimulated cells. In TNF-a stimulated cells MCP-1 protein synthesis was decreased by 35 ± 11% after siRNA transfection. Using a synthetic double-stranded siRNA, the TNF-a induced MCP-1 protein secretion could be successfully inhibited about 62.3 ± 10.3% in HEK 293 cells, indicating that the siRNA is functional in these cells to suppress chemokine expression. The siRNA approach targeting murine RANTES in Th1 cells and b-end3 cells revealed no inhibition of endogenous gene expression. Gene therapy approaches rely on efficient transfer of genes to the desired target cells. A wide variety of viral and nonviral vectors have been developed and evaluated for their efficiency of transduction, sustained expression of the transgene, and safety. Among them, lentiviruses have been widely used for gene therapy applications. In order to improve the delivery of TFOs or siRNAs into the target cells, cloning of the lentiviral transfer vector SEW, the production of lentiviral particles by transient transfection were performed with the aim to generate lentiviral vector-derived TFOs in further experiments. Here, Th1 cells were transduced with infectious lentiviral particles and transduction efficacy was measured. Transduction efficacy higher than 82% could be achieved using the lentiviral vector SEW, opening optimal possibilities for the TFO or siRNA approach.
(Non)retroflexivity of slavic affricates and its motivation : Evidence from polish and czech <č>

The goal of this paper is two-fold. First, it revises the common assumption that the affricate <č> denotes /t͡ʃ/ for all Slavic languages. On the basis of experimental results it is shown that Slavic <č> stands for two sounds: /t͡ʃ/ as e.g. in Czech and /ʈʂ/ as in Polish.
The second goal of the paper is to show that this difference is not accidental but it is motivated by perceptual relations among sibilants. In Polish, /t͡ʃ/ changed to /ʈʂ/ thus lowering its sibilant tonality and creating a better perceptual distance to /tɕ/, whereas in Czech /t͡ʃ/ did not turn to /ʈʂ/, as the former displayed sufficient perceptual distance to the only affricate present in the inventory, namely, the alveolar /t͡s/. Finally, an analysis of Czech and Polish affricate inventories is offered.
5-Lipoxygenase (5-LO) catalysis is positively regulated by Ca2+ ions and phospholipids that both act via the N-terminal C2-like domain of 5-LO. Previously, we have shown that 1-oleoyl-2-acetylglycerol (OAG) functions as an agonist for human polymorphonuclear leukocytes (PMNL) in stimulating 5-LO product formation. Here we have demonstrated that OAG directly stimulates 5-LO catalysis in vitro. In the absence of Ca2+ (chelated using EDTA), OAG strongly and concentration-dependently stimulated crude 5-LO in 100,000 x g supernatants as well as purified 5-LO enzyme from PMNL. Also, the monoglyceride 1-O-oleyl-rac-glycerol and 1,2-dioctanoyl-sn-glycerol were effective, whereas various phospholipids did not stimulate 5-LO. However, in the presence of Ca2+, OAG caused no stimulation of 5-LO. Also, phospholipids or cellular membranes abolished the effects of OAG. As found previously for Ca2+, OAG renders 5-LO activity resistant against inhibition by glutathione peroxidase activity, and this effect of OAG is reversed by phospholipids. Intriguingly, a 5-LO mutant lacking tryptophan residues (Trp-13, -75, and -102) important for the binding of the 5-LO C2-like domain to phospholipids was not stimulated by OAG. We conclude that OAG directly stimulates 5-LO by acting at a phospholipid binding site located within the C2-like domain.
An account of the bryological activities on the Azores is given. Three phases can be distinguished: one at the end of the 19. century, when first collections of non-bryologists were made, which resulted in the discovery of less than one third of the presently known species and the description of many so called endemics. A second phase in the thirties of the 20. century is characterized by an intensive collecting activity of some bryologists, which enormously raised the species numbers. In a third phase at the end of the 20. century, tourism as well as activities of local bryologists raised again the species numbers. Revisions of genera reduced the number of endemic species and revisions of herbarium specimens of doubtful records even reduced the species numbers of hepatics.
A bryophyte checklist of the Ecological Reserve of Gurjaú (08º10’00'’-08º15’00"S; 35º02’30"- 35º05’00"W), a remnant of the Atlantic Forest in the State of Pernambuco, has been compiled. The Reserve covers an area of approximately 1362 ha, including several forest fragments of diverse sizes and stages of conservation. Specimens collected during the years 2000 and 2001, and voucher specimens from the UFP Herbarium at the Federal University of Pernambuco (UFPE) were analyzed. Bryophytes were collected on live and dead trunks, rocks, leaves and soil. The studied bryoflora is composed of 53 species of liverworts, 37 species of mosses and one hornwort. Among the twenty-three registered families, Lejeuneaceae (41 spp.) was the most representative one with the highest generic and specific richness, which confirms its status as predominant in tropical rain forests. Fissidens (7 spp.), Lejeunea and Cheilolejeunea (6 spp.), and Calymperes (4 spp.) were the most representative genera. The studied bryoflora is predominantly composed of species of neotropical distribution, widely spread throughout Brazil. The presence of an endemic species of the Atlantic Forest, Vitalianthus bichlerianus (Pôrto & Grolle) Schust. & Giancotti, and some other exclusively Brazilian species, e.g., Riccardia regnellii (Ängstr.) Hell have been recorded from the site.
The Distrito Federal, one of the most important centers of plant endemism in central Brazil, is located in the center of Brazil, with an area of 5,814 km2 at coordinates 15o30’ S, 16o03’W. Cerrado vegetation covers the region. The main city in the Distrito Federal is Brasília, the capital of Brazil with a population of 2.5 million. In the last two decades the anthropogenic action has reduced the original plant cover drastically. In this paper a checklist of the bryophytes (Marchantiophyta, Bryophyta and Anthocerotophyta) is presented. A total of 176 species are listed, 114 among Bryophyta, 61 among Marchantiophyta and 1 in the Anthocerotophyta. The families best represented are the Bryaceae and Sematophyllaceae (Bryophyta), and the Jubulaceae and Lejeuneaceae (Marchantiophya).
This paper proves correctness of Nöcker's method of strictness analysis, implemented in the Clean compiler, which is an effective way for strictness analysis in lazy functional languages based on their operational semantics. We improve upon the work of Clark, Hankin and Hunt did on the correctness of the abstract reduction rules. Our method fully considers the cycle detection rules, which are the main strength of Nöcker's strictness analysis. Our algorithm SAL is a reformulation of Nöcker's strictness analysis algorithm in a higher-order call-by-need lambda-calculus with case, constructors, letrec, and seq, extended by set constants like Top or Inf, denoting sets of expressions. It is also possible to define new set constants by recursive equations with a greatest fixpoint semantics. The operational semantics is a small-step semantics. Equality of expressions is defined by a contextual semantics that observes termination of expressions. Basically, SAL is a non-termination checker. The proof of its correctness and hence of Nöcker's strictness analysis is based mainly on an exact analysis of the lengths of normal order reduction sequences. The main measure being the number of 'essential' reductions in a normal order reduction sequence. Our tools and results provide new insights into call-by-need lambda-calculi, the role of sharing in functional programming languages, and into strictness analysis in general. The correctness result provides a foundation for Nöcker's strictness analysis in Clean, and also for its use in Haskell.
Multicomponent Tree Adjoining Grammars (MCTAG) is a formalism that has been shown to be useful for many natural language applications. The definition of MCTAG however is problematic since it refers to the process of the derivation itself: a simultaneity constraint must be respected concerning the way the members of the elementary tree sets are added. Looking only at the result of a derivation (i.e., the derived tree and the derivation tree), this simultaneity is no longer visible and therefore cannot be checked. I.e., this way of characterizing MCTAG does not allow to abstract away from the concrete order of derivation. Therefore, in this paper, we propose an alternative definition of MCTAG that characterizes the trees in the tree language of an MCTAG via the properties of the derivation trees the MCTAG licences.
Artificial drainage of agricultural land, for example with ditches or drainage tubes, is used to avoid water logging and to manage high groundwater tables. Among other impacts it influences the nutrient balances by increasing leaching losses and by decreasing denitrification. To simulate terrestrial transport of nitrogen on the global scale, a digital global map of artificially drained agricultural areas was developed. The map depicts the percentage of each 5’ by 5’ grid cell that is equipped for artificial drainage. Information on artificial drainage in countries or sub-national units was mainly derived from international inventories. Distribution to grid cells was based, for most countries, on the "Global Croplands Dataset" of Ramankutty et al. (1998) and the "Digital Global Map of Irrigation Areas" of Siebert et al. (2005). For some European countries the CORINE land cover dataset was used instead of the both datasets mentioned above. Maps with outlines of artificially drained areas were available for 6 countries. The global drainage area on the map is 167 Mio hectares. For only 11 out of the 116 countries with information on artificial drainage areas, sub-national information could be taken into account. Due to this coarse spatial resolution of the data sources, we recommended to use the map of artificially drained areas only for continental to global scale assessments. This documentation describes the dataset, the data sources and the map generation, and it discusses the data uncertainty.
The Land and Water Development Division of the Food and Agriculture Organization of the United Nations and the Johann Wolfgang Goethe University, Frankfurt am Main, Germany, are cooperating in the development of a global irrigation-mapping facility. This report describes an update of the Digital Global Map of Irrigated Areas for the continent of Asia. For this update, an inventory of subnational irrigation statistics for the continent was compiled. The reference year for the statistics is 2000. Adding up the irrigated areas per country as documented in the report gives a total of 188.5 million ha for the entire continent. The total number of subnational units used in the inventory is 4 428. In order to distribute the irrigation statistics per subnational unit, digital spatial data layers and printed maps were used. Irrigation maps were derived from project reports, irrigation subsector studies, and books related to irrigation and drainage. These maps were digitized and compared with satellite images of many regions. In areas without spatial information on irrigated areas, additional information was used to locate areas where irrigation is likely, such as land-cover and land-use maps that indicate agricultural areas or areas with crops that are usually grown under irrigation. The study offers a discourse-based account of the Spanish copula forms ser and estar, which are generally considered to be lexical exponents of the stage-level/individual-level contrast. It argues against the popular view that the distinction between SLPs and ILPs rests on a fundamental cognitive division of the world that is reflected in the grammar. As it happens, conceptual oppositions like “temporary vs. permanent” or “arbitrary vs. essential“ provide only a preference for the interpretation of estar and ser. In addition, the evidence for an SLP/ILP impact on the grammar turns out to be far less conclusive than is currently assumed. The study argues against event-based accounts of the ser/estar contrast in particular, showing that ser and estar pattern alike in failing all of the standard eventuality tests. The discourse-based account proposed instead assumes that ser and estar both display the same lexical semantics (which is identical to the semantics of English be, German sein, etc.); estar differs from ser only in presupposing a relation to a specific discourse situation. By using estar a speaker restricts his or her claim to a specific discourse situation, whereas by using ser, the speaker makes no such restriction. The preference for interpreting estar predications as denoting temporary properties and ser predications as denoting permanent properties follows from economy principles driving the pragmatic legitimation of estars discourse dependence. The analysis proposed in this paper can also account for the observation that ser predications do not give rise to thetic judgements. The proposal is couched in terms of the framework of DRT.
We selectively survey, unify and extend the literature on realized volatility of financial asset returns. Rather than focusing exclusively on characterizing the properties of realized volatility, we progress by examining economically interesting functions of realized volatility, namely realized betas for equity portfolios, relating them both to their underlying realized variance and covariance parts and to underlying macroeconomic fundamentals.
A fresh look at the understanding of charity : with special reference to the present Indian practice

The discussion of this paper is divided in two parts: Present understanding of Charity and a fresh look at the understanding of Charity particularly with a reference to the present Indian practice. As the major religious and theological perspectives specially from the Christian and the Islamic point of views are being dealt by other presentations in this seminar, therefore, the discussion here is limited only first, to the dictionary based linguistic meaning of Charity as understood in three main English speaking contexts and then (second), the Charity as practised in the Indian context. The new and revised deluxe edition of the Webster’s Encyclopaedic Unbridged Dictionary of the English Language has given the following meanings of Charity: 1)charitable actions, as almsgiving or performing other benevolent actions of any sort for the needy with no expectation of material reward: to devote ones life to charity, 2)something given to a person or persons in need; aims: she asked for work not charity, 3) a charitable act or work 4) a charitable fund, foundation, or institution: He left his estate to one of his charities, 5) benevolent feeling, esp. towards those in need or in disfavour: she looked so poor that we fed her out of charity, and 6) Christian love; agape 1 Cor. 13. The Chamber English Dictionary, the meanings of Charity gives as: universal love (N.T.): the disposition to think favourably of others, and do them good almsgiving: a usu. non profit-making foundation, institution, or cause, devoted to caring for those in need of help etc. According to Concise Oxford Dictionary Charity means: an organisation set up to provide help and raise money for those in need, the voluntary giving of money those in need, tolerance in judging others and love of humankind, typically in Christian context. These three set of meanings of Charity, represent the three English regions or contexts: American (Webster), Scottish (Chamber) and English (Oxford). The common important element in all for these three is, the Christian understanding of Charity, because all the three directly have referred to the Biblical usages of Charity in some forms. Here for the discussion of this paper, one can also add that even these usages are limited to the contextual understanding of the English world. Because language is also considered as an vehicle of a culture. But it is true that even the English speaking persons from the non-English world particularly of the Southern countries, (where English rulers have ruled in the past), understand the meaning of Charity more or less in the above sense only. ...
I derive a general effective theory for hot and/or dense quark matter. After introducing general projection operators for hard and soft quark and gluon degrees of freedom, I explicitly compute the functional integral for the hard quark and gluon modes in the QCD partition function. Upon appropriate choices for the projection operators one recovers various well-known effective theories such as the Hard Thermal Loop/ Hard Dense Loop Effective Theories as well as the High Density Effective Theory by Hong and Schaefer. I then apply the effective theory to cold and dense quark matter and show how it can be utilized to simplify the weak-coupling solution of the color-superconducting gap equation. In general, one considers as relevant quark degrees of freedom those within a thin layer of width 2 Lambda_q around the Fermi surface and as relevant gluon degrees of freedom those with 3-momenta less than Lambda_gl. It turns out that it is necessary to choose Lambda_q << Lambda_gl, i.e., scattering of quarks along the Fermi surface is the dominant process. Moreover, this special choice of the two cutoff parameters Lambda_q and Lambda_gl facilitates the power-counting of the numerous contributions in the gap-equation. In addition, it is demonstrated that both the energy and the momentum dependence of the gap function has to be treated self-consistently in order to determine the imaginary part of the gap function. For quarks close to the Fermi surface the imaginary part is calculated explicitly and shown to be of sub-subleading order in the gap equation.
LIN-2/7 (L27) domains are protein interaction modules that preferentially hetero-oligomerize, a property critical for their function in directing specific assembly of supramolecular signaling complexes at synapses and other polarized cell-cell junctions. We have solved the solution structure of the heterodimer composed of the L27 domains from LIN-2 and LIN-7. Comparison of this structure with other L27 domain structures has allowed us to formulate a general model for why most L27 domains form an obligate heterodimer complex. L27 domains can be divided in two types (A and B), with each heterodimer comprising an A/B pair. We have identified two keystone positions that play a central role in discrimination. The residues at these positions are energetically acceptable in the context of an A/B heterodimer, but would lead to packing defects or electrostatic repulsion in the context of A/A and B/B homodimers. As predicted by the model, mutations of keystone residues stabilize normally strongly disfavored homodimers. Thus, L27 domains are specifically optimized to avoid homodimeric interactions.
A guide to bryological hotspots in Europe : [Part 2: The Rur Valley, Eifel Mountains, Germany]

The Rur valley is situated in the extreme west of Germany in the western part of the Eifel Mountains, close to the Belgian border. The Rur valley harbours several interesting, mainly bryopyhtes. Most of the interesting species are aquatic species. Very obvious is the oceanic element, which is represented by species such as Platyhypnidium lusitanicum, Hyocomium arnoricum, and Isothecium holtii.
In the last years, much effort went into the design of robust anaphor resolution algorithms. Many algorithms are based on antecedent filtering and preference strategies that are manually designed. Along a different line of research, corpus-based approaches have been investigated that employ machine-learning techniques for deriving strategies automatically. Since the knowledge-engineering effort for designing and optimizing the strategies is reduced, the latter approaches are considered particularly attractive. Since, however, the hand-coding of robust antecedent filtering strategies such as syntactic disjoint reference and agreement in person, number, and gender constitutes a once-for-all effort, the question arises whether at all they should be derived automatically. In this paper, it is investigated what might be gained by combining the best of two worlds: designing the universally valid antecedent filtering strategies manually, in a once-for-all fashion, and deriving the (potentially genre-specific) antecedent selection strategies automatically by applying machine-learning techniques. An anaphor resolution system ROSANA-ML, which follows this paradigm, is designed and implemented. Through a series of formal evaluations, it is shown that, while exhibiting additional advantages, ROSANAML reaches a performance level that compares with the performance of its manually designed ancestor ROSANA.
This paper contributes to the understanding of vocal folds oscillation during phonation. In order to test theoretical models of phonation, a new experimental set-up using a deformable vocal folds replica is presented. The replica is shown to be able to produce self sustained oscillations under controlled experimental conditions. Therefore different parameters, such as those related to elasticity, to acoustical coupling or to the subglottal pressure can be quantitatively studied. In this work we focused on the oscillation fundamental frequency and the upstream pressure in order to start (on-set threshold) either end (off-set threshold) oscillations in presence of a downstream acoustical resonator. As an example, it is shown how this data can be used in order to test the theoretical predictions of a simple one-mass model.
Dialogue has become a fashionable word in the theological circles for quite some time now. However, there is a need to review what has been achieved so far. If it is significant, we should then review how much religious tension has been reduced so far. If it is not much, why has there been no progress. In this note I will deal with the issues relating to Hindu-Christian dialogue. I am using Christianity only as a reference point, and the issues raised do have a wider context as well. As far as Hindu-Christian dialogue is concerned, I am of the opinion that there has been hardly any progress all these years. Many academics and theologians have been involved in the exercise so far. The whole literature, over a long period of time, seems to follow a familiar pattern – a discussion on the theory of the dialogue, what should be included in a dialogue, who should and should not be involved in a dialogue, and ends with a lament that there is so very little progress. The problem, according to me, is that the dialogue does not even consider a need to discuss what is the basic difference between Hinduism and Christianity, and an inquiry into whether these come in the way of communal harmony. A dialogue is really not necessary if we are to discuss only what is similar between the two systems. ...
In this dissertation a non-deterministic lambda-calculus with call-by-need evaluation is treated. Call-by-need means that subexpressions are evaluated at most once and only if their value must be known to compute the overall result. Also called "sharing", this technique is inevitable for an efficient implementation. In the lambda-ND calculus of chapter 3 sharing is represented explicitely by a let-construct. Above, the calculus has function application, lambda abstractions, sequential evaluation and pick for non-deterministic choice. Non-deterministic lambda calculi play a major role as a theoretical foundation for concurrent processes or side-effected input/output. In this work, non-determinism additionally makes visible when sharing is broken. Based on the bisimulation method this work develops a notion of equality which respects sharing. Using bisimulation to establish contextual equivalence requires substitutivity within contexts, i.e., the ability to "replace equals by equals" within every program or term. This property is called congruence or precongruence if it applies to a preorder. The open similarity of chapter 4 represents a new concept, insofar that the usual definition of a bisimulation is impossible in the lambda-ND calculus. So in section 3.2 a further calculus lambda-Approx has to be defined. Section 3.3 contains the proof of the so-called Approximation Theorem which states that the evaluation in lambda-ND and lambda-Approx agrees. The foundation for the non-trivial precongruence proof is set out in chapter 2 where the trailblazing method of Howe is extended to be capable with sharing. By the use of this (extended) method, the Precongruence Theorem proves open similarity to be a precongruence, involving the so-called precongruence candidate relation. Joining with the Approximation Theorem we obtain the Main Theorem which says that open similarity of the lambda-Approx calculus is contained within the contextual preorder of the lambda-ND calculus. However, this inclusion is strict, a property whose non-trivial proof involves the notion of syntactic continuity. Finally, chapter 6 discusses possible extensions of the base calculus such as recursive bindings or case and constructors. As a fundamental study the calculus lambda-ND provides neither of these concepts, since it was intentionally designed to keep the proofs as simple as possible. Section 6.1 illustrates that the addition case and constructors could be accomplished without big hurdles. However, recursive bindings cannot be represented simply by a fixed point combinator like Y, thus further investigations are necessary.
In many languages, a passive-like meaning may be obtained through a noncanonical passive construction. The get passive (1b) in English, the se faire passive (2b) in French and the kriegen passive (3b) in German represent typical manifestations. This squib focuses on the behavior of the get-passive in English and discusses a number of restrictions associated with it as well as the status of get.
The 5'-terminal cloverleaf (CL)-like RNA structures are essential for the initiation of positive- and negative-strand RNA synthesis of entero- and rhinoviruses. SLD is the cognate RNA ligand of the viral proteinase 3C (3Cpro), which is an indispensable component of the viral replication initiation complex. The structure of an 18mer RNA representing the apical stem and the cGUUAg D-loop of SLD from the first 5'-CL of BEV1 was determined in solution to a root-mean-square deviation (r.m.s.d.) (all heavy atoms) of 0.59 A (PDB 1Z30). The first (antiG) and last (synA) nucleotide of the D-loop forms a novel ‘pseudo base pair’ without direct hydrogen bonds. The backbone conformation and the base-stacking pattern of the cGUUAg-loop, however, are highly similar to that of the coxsackieviral uCACGg D-loop (PDB 1RFR) and of the stable cUUCGg tetraloop (PDB 1F7Y) but surprisingly dissimilar to the structure of a cGUAAg stable tetraloop (PDB 1MSY), even though the cGUUAg BEV D-loop and the cGUAAg tetraloop differ by 1 nt only. Together with the presented binding data, these findings provide independent experimental evidence for our model [O. Ohlenschläger, J. Wöhnert, E. Bucci, S. Seitz, S. Häfner, R. Ramachandran, R. Zell and M. Görlach (2004) Structure, 12, 237–248] that the proteinase 3Cpro recognizes structure rather than sequence.
A preliminary annotated checklist of the Chrysomelidae of the Baja California peninsula, Mexico is presented based on literature records and the examination of approximately 16,000 chrysomelid specimens. Seven subfamilies and 218 described species have been identified. Ninty-one species are recognized from the Baja California peninsula for the first time. Twenty-nine species are listed as endemic. An additional 128 morphospecies have been identified as undescribed or not yet associated with described species of mainland Mexico or the United States. Adult host plant associations have been identified for approximately 120 species or 35% of the leaf beetle fauna of the region. Each of the 218 described species is associated with the eight plant communities as outlined by Wiggins (1980). Two dominant subfamilies comprise 76% of the leaf beetle fauna: Galerucinae, 87 species (40%) and Cryptocephalinae, 78 species (36%). The following new combinations are proposed: Neolema californica (Heinze, 1927), transferred from Lema Fabricius, 1798; Pseudoluperus histrio (Horn, 1895), transferred from Keitheatus Wilcox, 1965, and Diachus peninsularis (Schaeffer, 1906), transferred from Triachus J. L. LeConte, 1880. Lema peninsulae Crotch, 1873, is removed from synonymy with L. balteata J. L. LeConte, 1884 and reinstated as a valid species. A replacement name is proposed: Longitarsus bajaensis Andrews and Gilbert for Longitarsus bicolor Horn, 1894.
Under a conventional policy rule, a central bank adjusts its policy rate linearly according to the gap between inflation and its target, and the gap between output and its potential. Under "the opportunistic approach to disinflation" a central bank controls inflation aggressively when inflation is far from its target, but concentrates more on output stabilization when inflation is close to its target, allowing supply shocks and unforeseen fluctuations in aggregate demand to move inflation within a certain band. We use stochastic simulations of a small-scale rational expectations model to contrast the behavior of output and inflation under opportunistic and linear rules. Klassifikation: E31, E52, E58, E61. Parkella Chickering 1946 = Metacyrba F. O. The six valid described species of Metacyrba are diagnosed and re-illustrated to show previously unrecognized genitalic differences. Metacyrba similis Banks 1904 is resurrected as a subspecies, becoming Metacyrba taeniola similis Banks 1904, n. status. The female of Metacyrba pictipes Banks 1903 is described for the first time. Metacyrba arizonensis Barnes 1958 = Platycryptus arizonensis (Barnes 1958), n. comb., and Marpissa magna (Peckham & Peckham 1894) = Platycryptus magnus (Peckham & Peckham 1894), n. comb. Platycryptus broadwayi (Peckham & Peckham 1894) = Platycryptus magnus (Peckham & Peckham 1894), n. syn. [lectotypes and paralectotypes are designated for both names]. Metacyrba nigrosecta (Mello-Leitão 1945) = Balmaceda nigrosecta Mello-Leitão 1945, comb. restored. The genera Balmaceda Peckham & Peckham 1894, Metacyrba, and Platycryptus Hill 1979 are compared morphologically among themselves and to Breda Peckham & Peckham 1894 and Fuentes Peckham & Peckham 1894. The distributions of Balmaceda picta Peckham & Peckham 1894 and Metacyrba species are updated. Marpissa melanura F.O.P.-Cambridge 1901 is resurrected; it is not a synonym of Marpissa minor F.O.P.-Cambridge 1901 nor Platycryptus californicus (Peckham & Peckham 1888).
The higher classification of the Nocluoidea (Oenosandridae, Doidae, Notodontidae, Strepsimanidae, Nolidae, lymanlriidae, Arctiidae, Erebidae, Micronoctuidae, and Noctuidae) is reviewed from the perspective of the classification proposed by KITCHING and RAWLINS (1998). Several taxa are reinstated, described as new, synonymised, or redescribed. Some characters that have been inadequately described, poorly understood, or misinterpreted, are redescribed and discussed. One family, two subfamilies, four tribes, and three subtribes are proposed as new. Available family-group names of Noctuoidea are listed In an appendix.
It has been shown that visual cues play a crucial role in the perception of vowels and consonants. Conflicting consonantal stimuli presented in the visual and auditory modalities can even result in the emergence of a third perceptual unit (McGurk effect). From a developmental point of view, several studies report that newborns can associate the image of a face uttering a given vowel to the auditory signal corresponding to this vowel; visual cues are thus used by the newborns. Despite the large number of studies carried out with adult speakers and newborns, very little work has been conducted with preschool-aged children. This contribution is aimed at describing the use of auditory and visual cues by 4 and 5-year-old French Canadian speakers, compared to adult speakers, in the identification of voiced consonants. Audiovisual recordings of a French Canadian speaker uttering the sequences [aba], [ada], [aga], [ava], [ibi], [idi], [igi], [ivi] have been carried out. The acoustic and visual signals have been extracted and analysed so that conflicting and non-conflicting stimuli, between the two modalities, were obtained. The resulting stimuli were presented as a perceptual test to eight 4 and 5-year-old French Canadian speakers and ten adults in three conditions: visual-only, auditory-only, and audiovisual. Results show that, even though the visual cues have a significant effect on the identification of the stimuli for adults and children, children are less sensitive to visual cues in the audiovisual condition. Such results shed light on the role of multimodal perception in the emergence and the refinement of the phonological system in children.
A taxonomic revision of the genus Syntrichia Brid. (Pottiaceae, Musci) in the Mediterranean Region and Macaronesia has been carried out, thus contributing to knowledge of its distinguisliing morphological characters, geographic distribution and nomenclature. Some 3000 specimens, including the most of type material, were studied. An identification key, morphological descriptions, photographs and numerous observations on taxonomic and nomenclatural problems of the 23 taxa accepted in the study area, are provided. New records for some countries of the Mediterranean basin and Macaronesia are given. Five new Synonyms are proposed. The designations of 11 new lectotypes are included and the name Syntrichia aciphylla var. calva J.J. Amann is excluded from the genus Syntrichia.
A visual articulatory model and its application to therapy
of speech disorders : a pilot study

A visual articulatory model based on static MRI-data of isolated sounds and its application in therapy of speech disorders is described. The model is capable of generating video sequences of articulatory movements or still images of articulatory target positions within the midsagittal plane. On the basis of this model (1) a visual stimulation technique for the therapy of patients suffering from speech disorders and (2) a rating test for visual recognition of speech movements was developed. Results indicate that patients produce recognition rates above level of chance already without any training and that patients are capable of increasing their recognition rate over the time course of therapy significantly.
The main subject of this survey are Belyi functions and dessins d'enfants on Riemann surfaces. Dessins are certain bipartite graphs on 2-mainfolds defining there are conformal and even an algebraic structure. In principle, all deeper properties of the resulting Riemann surfaces or algebraic curves should be encoded in these dessins, but the decoding turns out to be difficult and leads to many open problems. We emphasize arithmetical aspects like Galois actions, the relation to the ABC theorem in function filds and arithemtic questions in uniformization theory of algebraic curves defined over number fields.
Mitochondial NADH:ubiquinone oxidoreductase (complex I) the largest multiprotein enzyme of the respiratory chain, catalyses the transfer of two electrons from NADH to ubiquinone, coupled to the translocation of four protons across the membrane. In addition to the 14 strictly conserved central subunits it contains a variable number of accessory subunits. At present, the best characterized enzyme is complex I from bovine heart with a molecular mass of about 980 kDa and 32 accessory proteins. In this study, the subunit composition of mitochondrial complex I from the aerobic yeast Y. lipolytica has been analysed by a combination of proteomic and genomic approaches. The sequences of 37 complex I subunits were identified. The sum of their individual molecular masses (about 930 kDa) was consistent with the native molecular weight of approximately 900 kDa for Y. lipolytica complex I obtained by BN-PAGE. A genomic analysis with Y. lipolytica and other eukaryotic databases to search for homologues of complex I subunits revealed 31 conserved proteins among the examined species. A novel protein named “X” was found in purified Y. lipolytica complex I by MALDI-MS. This protein exhibits homology to the thiosulfate sulfurtransferase enzyme referred to as rhodanese. The finding of a rhodanese-like protein in isolated complex I of Y. lipolytica allows to assume a special regulatory mechanism of complex I activity through control of the status of its iron-sulfur clusters. The second part of this study was aimed at investigating the possible role of one of these extra subunits, 39 kDa (NUEM) subunit which is related to the SDRs-enzyme family. The members of this family function in different redox and isomerization reactions and contain a conserved NAD(P)H-binding site. It was proposed that the 39 kDa subunit may be involved in a biosynthetic pathway, but the role of this subunit in complex I is unknown. In contrast to the situation in N. crassa, deletion of the 39 kDa encoding gene in Y. lipolytica led to the absence of fully assembled complex I. This result might indicate a different pathway of complex I assembly in both organisms. Several site-directed mutations were generated in the nucleotide binding motif. These had either no effect on enzyme activity and NADPH binding, or prevented complex I assembly. Mutations of arginine-65 that is located at the end of the second b-strand and responsible for selective interaction with the 2’-phosphate group of NADPH retained complex I activity in mitochondrial membranes but the affinity for the cofactor was markedly decreased. Purification of complex I from mutants resulted in decrease or loss of ubiquinone reductase activity. It is very likely that replacement of R65 not only led to a decrease in affinity for NADPH but also caused instability of the enzyme due to steric changes in the 39 kDa subunit. These data indicate that NADPH bound to the 39 kDa subunit (NUEM) is not essential for complex I activity, but probably involved in complex I assembly in Y. lipolytica.
The present article is a follow-up study of the investigation of labiodentals in German and Dutch by Hamann & Sennema (2005), where we looked at the perception of the Dutch labiodental three-way contrast by German listeners without any knowledge of Dutch and German learners of Dutch. The results of this previous study suggested that the German voiced labiodental fricative /v/ is perceptually closer to the Dutch approximant /ʋ/ than to the corresponding Dutch voiced labiodental fricative /v/. These perceptual indications are attested by the acoustic findings in the present study. German /v/ has a similar harmonicity median and a similar centre of gravity to Dutch /ʋ/, but differs from Dutch /v/ in these parameters. With respect to the acoustic parameter of duration, German /v/ lies closer to the Dutch /v/ than to the Dutch /ʋ/.
Four speakers repeated 8 times 15 sentences containing 'pVp' syllables (V being /a/, /i/ and /u/). The 'pVp' syllables were located in final, penultimate and antepenultimate position relatively to the Intonational Phrase (IP) boundary. They were embedded in lexical words of 1-3 syllables and were either word-initial or word-final. Results show that the closer the vowel in word-final position is to the IP boundary, the longer the duration and the higher the fundamental frequency of the vowel; it is also characterised by larger lip opening gestures. The potential reduction or coarticulation of vowels in wordinitial position compared to their counterparts in word-final position is discussed.
