Von einem Absoluten zu einem Anderen : Celan und Blanchot: "Sprich als letzter, sag deinen Spruch"

Dieses kurze Zitat aus dem 1980 veröffentlichten Essay von Blanchot "Die Schrift des Desasters" ist eine der vielen Überlegungen des Autors über den Holocaust. Jene eingehende Betrachtung Blanchots über «das absolute Ereignis der Geschichte», die sein ganzes Werk durchzieht, hat ihn dazu geführt, die Literatur neu durchzudenken und seine eigene Antwort auf die berühmt gewordene Frage Adornos zu finden. Mit seiner kurzen Erzählung "Der Wahnsinn des Tages" hatte der Autor und Essayist schon die Frage nach der Möglichkeit gestellt, eine Erzählung nach der Erfahrung der Shoah zu schreiben, es ging dabei um eine Art Geständnis des Schriftstellers über die Unmöglichkeit, weiterhin zu dichten. Somit wird das Fragmentarische in seinen Büchern immer wichtiger; die Entstehung und die Entwicklung dieses neuen Schreibens hängt auch mit einem ständigen Dialog mit einem anderen Denken, mit anderen Autoren zusammen, die für sein Werk von großer Bedeutung sind, wie z. B. Paul Celan.
In dieser Arbeit sollte die Bindung von Tetrahydromethanopterinderivaten an zwei Enzyme des methanogenen, CO2-reduzierenden Energiestoffwechselweges strukturell charakterisiert werden. In jenem Stoffwechselweg verläuft die schrittweise Reduktion von CO2 über die Bindung an den C1-Carrier Tetrahydromethanopterin (H4MPT), ein Tetrahydrofolat-Analogon, welches unter anderem in methanogenen Archaeen zu finden ist. Die thermophilen bzw. hyperthermophilen Ursprungsorganismen der untersuchten Enzyme, Methanothermobacter marburgensis, Methanocaldococcus jannaschii und Methanopyrus kandleri, sind aufgrund ihrer Anpassung an extreme Habitate durch spezielle genomische, strukturelle und enzymatische Eigenschaften von strukturbiologischem Interesse. Beim ersten in dieser Arbeit untersuchten Enzym handelte es sich um den aus acht Untereinheiten bestehenden membrangebundenen N5-Methyl-H4MPT:Coenzym M-Methyltransferasekomplex (MtrA-H). Dieser katalysiert in einem zweistufigen Mechanismus den Methyltransfer von H4MPT zum Co(I) der prosthetischen Gruppe 5’-Hydroxybenzimidazolylcobamid (Vitamin B12a), um die Methylgruppe dann auf Coenzym M zu übertragen. Gleichzeitig findet ein der Energiekonservierung dienender vektorieller Natriumtransport über die Membran statt. Für den Mtr-Komplex aus M. marburgensis (670 kDa) lag bereits ein Protokoll zur Reinigung unter anaeroben Bedingungen vor. Dieses wurde im Rahmen dieser Arbeit verbessert, für die Isolierung und Reinigung unter aeroben Bedingungen vereinfacht und für die Erfordernisse der zur Strukturbestimmung verwendeten elektronenmikroskopischen Einzelpartikelmessung optimiert. Neben der Präparation des kompletten Komplexes MtrA-H wurde als Alternative die Präparation des Enzymkomplexes MtrA-G unter möglichst vollständiger Abtrennung der hydrophilsten Untereinheit MtrH gewählt. Mit der zu diesem Zweck entwickelten Methode konnte das Abdissoziieren von MtrH besser als im etablierten Protokoll kontrolliert und somit die Homogenität der Probe deutlich verbessert werden. Dies schafft zum einen die Vorraussetzungen für eine Kristallisation zur Röntgenstrukturanalyse, zum anderen war auch in bei der elektronenmikroskopischen Einzelpartikelmessung erkennbar, dass mit dem Mtr-Komplex ohne MtrH bessere Ergebnisse zu erzielen sind. Parallel zu den Untersuchungen am Gesamtkomplex sollten die den Cobamid-Cofaktor bindende Untereinheit MtrA sowie die H4MPT-bindende Untereinheit MtrH in für die Kristallisation und röntgenkristallographische Untersuchung ausreichender Menge und Qualität gereinigt werden. Hierfür wurden MtrA und MtrH aus oben genannten Organismen für die heterologe Expression in E. coli kloniert, die Expressionsbedingungen optimiert und Reinigungsprotokolle etabliert. Anschließend wurden die Untereinheiten umfangreichen Kristallisationsversuchen unterzogen. Die Untereinheit MtrA aus M. jannaschii konnte ohne die C-terminale Transmembranhelix als lösliches Protein in E. coli produziert und als Holoprotein bis zur Homogenität gereinigt werden. Bei M. kandleri MtrA gelang die Herstellung von geringen Mengen teilweise löslichen StrepII-Fusionsproteins ohne C-terminale Transmembranhelix in E. coli. Eine Produktion der Untereinheit MtrH in E. coli als lösliches Protein war bei keiner der in dieser Arbeit getesteten Varianten möglich. Mit dem in Einschlusskörperchen exprimierten Protein aus M. marburgensis wurde eine Reinigung und Rückfaltung versucht. Auch eine Co-Expression der Untereinheiten MtrA und MtrH, durch welche eine bessere Faltung und Löslichkeit erreicht werden sollte, war nur in Einschlusskörperchen möglich. Das zweite in dieser Arbeit untersuchte Enzym, die F420 abhängige N5,N10 Methylen-H4MPT-Dehydrogenase (Mtd), katalysiert den reversiblen, stereospezifischen Hydrid-Transfer zwischen reduziertem F420 (F420H2) und Methenyl-H4MPT+, welches hierbei zu Methylen-H4MPT reduziert wird. Die Reaktion verläuft über einen ternären Komplex bestehend aus Protein, Substrat (Methylen-H4MPT) und Cosubstrat (F420), welcher strukturell charakterisiert werden sollte. Das gereinigte, rekombinante Enzym aus M. kandleri wurde mit verschiedenen H4MPT- und F420-Derivaten co-kristallisiert, die Struktur des ternären Komplexes röntgenkristallographisch bestimmt und die Bindung von H4MPT und F420 analysiert. Methenyl-H4MPT+ und F420H2 sind in der in dieser Arbeit gelösten Kristallstruktur in katalytisch aktiver Konformation gebunden, jedoch kann bei einer Auflösung von 1,8 Å nicht beurteilt werden, ob Methylen-H4MPT und F420 oder Methenyl-H4MPT+ und F420H2 vorlagen. Ein Vergleich mit der Struktur von M. kandleri-Mtd (KMtd) ohne Substrat und Cosubstrat ergab nur äußerst geringe Abweichungen in der Proteinkonformation, sodass sich KMtd überraschenderweise als Beispiel für ein Enzym mit ungewöhnlich starrer, vorgegebener Bindetasche erwies.

We previously proposed that the dimeric cytochrome bc(1) complex exhibits half-of-the-sites reactivity for ubiquinol oxidation and rapid electron transfer between bc(1) monomers (Covian, R., Kleinschroth, T., Ludwig, B., and Trumpower, B. L. (2007) J. Biol. Chem. 282, 22289-22297). Here, we demonstrate the previously proposed half-of-the-sites reactivity and intermonomeric electron transfer by characterizing the kinetics of ubiquinol oxidation in the dimeric bc(1) complex from Paracoccus denitrificans that contains an inactivating Y147S mutation in one or both cytochrome b subunits. The enzyme with a Y147S mutation in one cytochrome b subunit was catalytically fully active, whereas the activity of the enzyme with a Y147S mutation in both cytochrome b subunits was only 10-16% of that of the enzyme with fully wild-type or heterodimeric cytochrome b subunits. Enzyme with one inactive cytochrome b subunit was also indistinguishable from the dimer with two wild-type cytochrome b subunits in rate and extent of reduction of cytochromes b and c(1) by ubiquinol under pre-steady-state conditions in the presence of antimycin. However, the enzyme with only one mutated cytochrome b subunit did not show the stimulation in the steady-state rate that was observed in the wild-type dimeric enzyme at low concentrations of antimycin, confirming that the half-of-the-sites reactivity for ubiquinol oxidation can be regulated in the wild-type dimer by binding of inhibitor to one ubiquinone reduction site.
We present a tractable model of the effects of nonfinancial risk on intertemporal choice. Our purpose is to provide a simple framework that can be adopted in fields like representative-agent macroeconomics, corporate finance, or political economy, where most modelers have chosen not to incorporate serious nonfinancial risk because available methods were too complex to yield transparent insights. Our model produces an intuitive analytical formula for target assets, and we show how to analyze transition dynamics using a familiar Ramsey-style phase diagram. Despite its starkness, our model captures most of the key implications of nonfinancial risk for intertemporal choice.
American households have received a triple dose of bad news since the beginning of the current recession: The greatest collapse in asset values since the Great Depression, a sharp tightening in credit availability, and a large increase in unemployment risk. We present measures of the size of these shocks and discuss what a benchmark theory says about their immediate and ultimate consequences. We then provide a forecast based on a simple empirical model that captures the effects of wealth shocks and unemployment fears. Our short-term forecast calls for somewhat weaker spending, and somewhat higher saving rates, than the Consensus survey of macroeconomic forecasters. Over the longer term, our best guess is that the personal saving rate will eventually approach the levels that preceded period of financial liberalization that began in the late 1970s. We model the motives for residents of a country to hold foreign assets, including the precautionary motive that has been omitted from much previous literature as intractable. Our model captures many of the principal insights from the existing specialized literature on the precautionary motive, deriving a convenient formula for the economy’s target value of assets. The target is the level of assets that balances impatience, prudence, risk, intertemporal substitution, and the rate of return. We use the model to shed light on two topical questions: The “upstream” flows of capital from developing countries to advanced countries, and the long-run impact of resorbing global financial imbalances
The budget constraint requires that, eventually, consumption must adjust fully to any permanent shock to income. Intuition suggests that, knowing this, optimizing agents will fully adjust their spending immediately upon experiencing a permanent shock. However, this paper shows that if consumers are impatient and are subject to transitory as well as permanent shocks, the optimal marginal propensity to consume out of permanent shocks (the MPCP) is strictly less than 1, because buffer stock savers have a target wealth-to-permanent-income ratio; a positive shock to permanent income moves the ratio below its target, temporarily boosting saving. This articles aims to present some of the main concepts of E. Husserl's phenomenology that can be applied to linguistic communication. The apprehension of those concepts is condition sine qua non for the use of the phenomenology as a matrix for research. The understanding of those concepts will serve as a work instrument in the field of applied linguistic.
War and Peace in Contemporary Eritrean Poetry focuses on Eritrean written poetry from roughly the last three decades of the twentieth century. The poems appear in the anthology Who Needs a Story? Contemporary Eritrean Poetry in Tigrinya, Tigre and Arabic from which a selection is offered here in their original scripts of Ge'ez or Arabic, and in English translation. Who Needs a Story? is the first anthology of contemporary poetry from Eritrea ever published, and War and Peace in Contemporary Eritrean Poetry is the first book on the subject. Therefore, the groundbreaking effort of the former warrants a discussion of its means of cultural production. All of the poets in Who Needs a Story? participated in the Eritrean struggle for independence (1961-91) as freedom fighters and/or as supporters in the Eritrean diaspora. Thus, contemporary Eritrean poetry divides itself between experiences of war and peace, although one can contain the other as well. War and Peace in Contemporary Eritrean Poetry also includes an extended analysis of one of Eritrea's most famous contemporary poets Reesom Haile, as an example of the kind of extended analysis that many of the poets of Who Needs a Story? should stimulate and, last but not least, a meditation on how the author, a non-native speaker, personally becomes involved in Eritrean poetry translation.
Seit dem Inkrafttreten des Investmentänderungsgesetzes zum 28.12.2007 steht der Investmentbranche als neue Gestaltungsform eines Investmentvehikels die fremdverwaltete Investmentaktiengesellschaft zur Verfügung. Die fremdverwaltete Investmentaktiengesellschaft benennt eine Kapitalanlagegesellschaft als Verwaltungsgesellschaft und überträgt ihr die allgemeine Verwaltungstätigkeit sowie die Anlage und Verwaltung ihrer Mittel. Der folgende Beitrag untersucht die Haftung der Verwaltungsgesellschaft gegenüber den Aktionären der fremdverwalteten Investmentaktiengesellschaft. Im Ergebnis wird ein gesetzliches Schuldverhältnis bejaht, für dessen Verletzung die Verwaltungsgesellschaft von den Aktionären der Investmentaktiengesellschaft gemäß §§ 280 Abs. 1, 249 ff. BGB auf Schadensersatz in Anspruch genommen werden kann.
Durch das MoMiG ist der durch das sog. Novemberurteil begründeten restriktiven Rechtsprechung des BGH zur grundsätzlichen Unzulässigkeit von Darlehen aus gebundenem Vermögen an Gesellschafter die Grundlage entzogen worden. In seiner MPS-Entscheidung vom 1. Dezember 2008 hat der BGH daraus die Konsequenzen für §§ 311 ff. AktG gezogen. Entscheidend ist nunmehr, ob der Rückerstattungsanspruch vollwertig ist oder ob ein konkretes Ausfallrisiko besteht. Der Beitrag analysiert die Implikationen der Entscheidung für die Vermögensbindung und das Recht des faktischen Konzerns. Der Schwerpunkt liegt dabei auf der Frage, wie die Kriterien der Vollwertigkeit bzw. des konkreten Ausfallrisikos zu konkretisieren sind.
Die Verbreitung und Vorkommen der im östlichen Ruhrgebiet, besonders in Dortmund und Umgegend, bisher aufgetretenen Vertreter der Gattung Liebesgras werden dargestellt. Insbesondere wird auf Eragrostis minor, das Kleine Liebesgras, eingegangen, welches schon längst nicht mehr das typische Gras von Bahnanlagen ist. Die jüngst erfolgte, scheinbar explosionsartige Ausbreitung des Japanischen Liebesgrases (Eragrostis multicaulis s. l.) wird näher beschrieben.
Mitochondria are essential for respiration and oxidative phosphorylation. Mitochondrial dysfunction due to aging processes is involved in pathologies and pathogenesis of a series of cardiovascular disorders. New results accumulate showing that the enzyme telomerase with its catalytic subunit telomerase reverse transcriptase (TERT) has a beneficial effect on heart functions. The benefit of short-term running of mice for heart function is dependent on TERT expression. TERT can translocate into the mitochondria and mitochondrial TERT (mtTERT) is protective against stress induced stimuli and binds to mitochondrial DNA (mtDNA). Because mtDNA is highly susceptible to damage produced by reactive oxygen species (ROS) which are generated in close proximity to the respiratory chain, the aim of this study was to determine the functions of mtTERT in vivo and in vitro. Therefore, mitochondria from hearts of adult, 2nd generation TERT-deficient mice (TERT -/-) and wt littermates were isolated and state 3 respiration was measured. Strikingly mitochondria from TERT -/- revealed a significantly lower state 3 respiration (TERTwt: 987 +/- 72 pmol/s*mg vs. TERT-/-: 774 +/- 38 pmol/s*mg, p < 0.05, n = 5). These results demonstrated that TERT -/- mice have a so far undiscovered heart phenotype. In contrast mitochondria isolated from liver tissues did not show any differences. To get further insights in the molecular mechanisms, we reduced endogenous TERT levels by shRNA and measured mitochondrial reactive oxygen species (mtROS). mtROS were increased after ablation of TERT (scrambled: 4.98 +/- 1.1% gated vs. shTERT: 2.03 +/- 0.7% gated, p < 0.05, n = 4). We next determined mtDNA deletions, which are caused by mtROS. Semiquantitative realtime PCR of mtDNA deletions revealed that mtTERT protects mtDNA from oxidative damage. To analyze whether mitochondrial integrity is required to protect from apoptosis, vectors with mitochondrially targeted TERT (mitoTERT) and wildtype TERT (wtTERT) were transfected and apoptosis was measured. mitoTERT showed the most prominent protective effect on H2O2 induced apoptosis. In conclusion, mtTERT has a protective role in mitochondria by importantly contributing to mtDNA integrity and thereby enhancing respiration capacity of the heart.
The seasonality of transport and mixing of air into the lowermost stratosphere (LMS) is studied using distributions of mean age of air and a mass balance approach, based on in-situ observations of SF6 and CO2 during the SPURT (Spurenstofftransport in der Tropopausenregion, trace gas transport in the tropopause region) aircraft campaigns. Combining the information of the mean age of air and the water vapour distributions we demonstrate that the tropospheric air transported into the LMS above the extratropical tropopause layer (ExTL) originates predominantly from the tropical tropopause layer (TTL). The concept of our mass balance is based on simultaneous measurements of the two passive tracers and the assumption that transport into the LMS can be described by age spectra which are superposition of two different modes. Based on this concept we conclude that the stratospheric influence on LMS composition is strongest in April with extreme values of the tropospheric fractions (alpha1) below 20% and that the strongest tropospheric signatures are found in October with alpha1 greater than 80%. Beyond the fractions, our mass balance concept allows us to calculate the associated transit times for transport of tropospheric air from the tropics into the LMS. The shortest transit times (<0.3 years) are derived for the summer, continuously increasing up to 0.8 years by the end of spring. These findings suggest that strong quasi-horizontal mixing across the weak subtropical jet from summer to mid of autumn and the considerably shorter residual transport time-scales within the lower branch of the Brewer-Dobson circulation in summer than in winter dominates the tropospheric influence in the LMS until the beginning of next year's summer.
CITIES (Chemistry and Industry for Teachers in European Schools) ist ein COMENIUS-Projekt, in dessen Rahmen Materialien für den Chemieunterricht erstellt und erprobt werden. Diese Materialien sollen Lehrkräften helfen, ihren Unterricht attraktiver zu gestalten, indem der Bezug sowohl zum Alltag und der Lebenswelt als auch zur chemischen Industrie aufgezeigt wird. Chemie des Alltags Nicht immer besteht die Zeit, im Unterricht ein zusammenhängendes größeres Projekt zu bearbeiten. Experimente, die chemische Hintergründe der Funktion von Produkten oder von Vorgängen aus der Alltags- und Lebenswelt der Schülerinnen und Schüler aufzeigen, können aber vielfach in den Unterricht integriert werden. Im dritten Bereich des Moduls wurde eine Auswahl solcher Experimente zusammengestellt. Im einzelnen finden sich Versuche zu folgenden Themen: 1. Bestimmung der Dicke der Aluminiumschicht einer Verpackungsfolie 2. Herstellen von Formaldhyd-Harnstoff-Leim und Nachweis dieses Leims in Spanplatten und Linoleum 3. Polyurethan auf der Basis von Ricinusöl 4. Nylon durch Grenzflächen-Polykondensation 5. Ascorbinsäure als Reduktionsmittel und Nachweis von Ascorbinsäure in Gemüse 6. Nachweis reduzierender Zucker 7. Nachweis von Proteinen 8. Herstellung von Biodiesel 9. Fluoreszenz von Pflanzenfarbstoffen
Seit 2003 wird die Krankenhausvergütung in Deutschland schrittweise auf ein neues, leistungsorientiertes System auf Basis von Fallpauschalen (diagnosis related groups – DRG) umgestellt. Die Über gangsphase, in der gesonderte gesetzliche Regelungen gegolten hatten, sollte Ende 2008 auslaufen, weshalb der gesetzlichen Rahmen für die Nachkonvergenzzeit neu geregelt werden musste. Neben diesen technischen Regelungen und einem einheitlichen Bundesbasisfallwert standen auch die Reform der Investitionsfinanzierung, der weitere Ausbau des Wettbewerbes und die schlechte Finanzsituation der Krankenhäuser auf der Agenda des Krankenhausfinanzierungsreformgesetzes (KHRG). Aufgrund der heterogenen Interessenslagen von Bund und Ländern, gestaltete sich auch bei diesem Reformgesetz die Kompromisssuche schwierig. Herausgekommen ist ein Gesetz, dass die Reformpolitik der letzten Jahre weiterführt, die Aufgabe der zukünftigen Sicherstellung der bedarfsgerechten Versorgung aber nicht löst und die problematischen Auswirkungen des DRG-Systems negiert. Das Diskussionspapier zeichnet die wichtigsten Schritte des Gesetzgebungsprozesses nach, präsentiert und diskutiert die wichtigsten Inhalte des Gesetzes und ordnet das KHRG in den Reformkontext der letzten Jahre ein.
To understand the rapid rate of change in global biodiversity, it is necessary to analyse the present condition of ecosystems and to elucidate relationships of species to their environment. The BIOKLIM Project (Biodiversity and Climate Change Project) is intended to close this gap in our knowledge of montane and high montane forests of Central European low mountain ranges, one of the most threatened mixed montane systems worldwide. The Bavarian Forest National Park is characterised by its altitude range of ca. 800 m and a strongly developed gradient of forest structure. Relicts of old growth forests (areas of former local nature reserves) and dead stands, mostly killed by bark beetles, are accompanied by widely varying levels of woody debris and light. The gradients comprise a wide range of abiotic and forest structure factors, making the study area well suited for a multidisciplinary investigation of biodiversity. Unconstrained ordination (CA) of six taxa (vascular plants, wood inhabiting fungi, birds, carabids, spiders and molluscs) indicate the altitudinal gradient to be the main driver for distribution patterns of species assemblages. Objectives, structure, study design and data sampling of the BIOKLIM Project are described in detail. We set up 293 sampling plots along four main straight transects following the altitudinal gradient. All abiotic and stand structure data regarded as relevant are available for each plot. Vascular plants, wood inhabiting fungi and birds were sampled or mapped on all 293 plots. For the other 22 investigated taxa we used subsamples pre-stratified according to the sampling methods. The necessity of dealing with spatial autocorrelation, arising from sampling along linear transects, is described. Finally, study approach of our biodiversity project is compared with others involving altitudinal gradients. Worldwide, only a few multidisciplinary biodiversity studies have been previously conducted on long altitudinal gradients. However, in most cases sampling techniques were similar to ours, which allows comparison of results between continents. Keywords: Climate Change, Biodiversity, species-environment relationships
