Within the visual cortex, it has been proposed that interhemispheric interactions serve to re-establish the continuity of the visual field across its vertical meridian (VM) by mechanisms similar to those used by intrinsic connections within a hemisphere. However, other specific functions of transcallosal projections have also been proposed, including contributing to disparity tuning and depth perception. Here, we consider whether interhemispheric connections modulate specific response properties, orientation and direction selectivity, of neurons in areas 17 and 18 of the ferret by combining reversible thermal deactivation in one hemisphere with optical imaging of intrinsic signals and single-cell electrophysiology in the other hemisphere. We found interhemispheric influences on both the strength and specificity of the responses to stimulus orientation and direction of motion, predominantly at the VM. However, neurons and domains preferring cardinal contours, in particular vertical contours, seem to receive stronger interhemispheric input than others. This finding is compatible with interhemispheric connections being involved in horizontal disparity tuning. In conclusion, our results support the view that interhemispheric interactions mainly perform integrative functions similar to those of connections intrinsic to one hemisphere. Bare incrusted soils are a degradation stage often encountered in the Sahel zone. Our study documents the success of restoration (= regreening) experiments using deep ploughing in an experimental site south of Gorom-Gorom in the Oudalan province of Burkina Faso. We used phytosociological relevés and maximum likelihood classifications of digital photography to analyze changes in vegetation. Plant cover in treated plots was found to be about 20 times higher than in control plots, mean species richness more than twice as high. Therefore, this promising restoration method should be tested also in other Sahelian regions. Our approach to combine phytosociological relevés and maximum likelihood classifications of digital photography proved to be very useful.
Morphin-6-Glukuronid (M6G) ist ein aktiver Metabolit von Morphin. Er ist mitverantwortlich für die analgetische und toxische Wirkung nach einer Morphinverabreichung. M6G wird aktuell in klinischen Phase-III-Studien getestet und möglicherweise zukünftig als ein alternatives Schmerzmittel zu Morphin zur Behandlung postoperativer Schmerzen eingeführt. Seine zentralnervösen Opioideffekte könnten sich aufgrund einer erhöhten Konzentration im zentralen Nervensystem (ZNS) als Folge einer erhöhten M6G Blutkonzentration und/ oder einem erhöhten ZNS Transport an der Blut-Hirn-Schranke verstärken. M6G scheint Substrat zahlreicher transmembranöser Transporter zu sein, die u.a. an der Blut-Hirn-Schranke, Niere oder Leber lokalisiert sind. An der Blut-Hirn-Schranke pumpen diese Transporter M6G aktiv aus dem ZNS zurück in das Blut. Die Hemmung der Transporteraktivität könnte somit eine potentielle Ursache einer erhöhten ZNS Konzentration von M6G mit der Folge vermehrter Nebenwirkungen unter Morphin bzw. Morphin-6-Glukuronid Administration sein. Wir untersuchten die Auswirkungen einer pharmakologischen Hemmung der transmembranösen Transporter P-Glykoprotein, sowie den Probenecid sensitiven Transportern multidrug resistance proteins (MRPs), organic anion transporters (OATs) und organic anion transporter polypeptides (OATPs) auf die Rückenmarkkonzentration und antinozizeptiven Effekte von M6G in Ratten. PSC 833 bzw. Probenecid wurden als Inhibitoren von P-Glykoprotein bzw. Probenecid sensitiven Transportern verwendet. Die Rückenmarkkonzentration von M6G wurde direkt mittels in vivo Mikrodialyse durch eine transversal in das Hinterhorn des Rückenmarks implantierten Sonde gemessen. 20 Ratten erhielten M6G als intravenöse Infusion für 8 Stunden, jeweils 5 von diesen bekamen zusätzlich eine Infusion mit PSC 833 oder Probenecid. Die antinozizeptiven Effekte von M6G wurden durch den Formalinschmerztest während der 8. Stunde der M6G Infusion in weiteren Verhaltensexperimenten bestimmt. Dabei wurde die Dosisabhängigkeit der antinozizeptiven Effekte von M6G mit oder ohne PSC 833 bzw. Probenecid im Vergleich mit PSC 833 alleine, Probenecid alleine und Placebo untersucht. In den Mikrodialyseexperimenten wurde unter P-Glykoprotein Hemmung mit PSC 833 eine nahezu dreifach höhere M6G Rückenmarkkonzentration gemessen, während die Blutplasmakonzentration nur um den Faktor 1.2 erhöht war. Die Folge war eine Verdoppelung des Konzentrationsverhältnis Rückenmark zu Blutplasma (von 0.08 ± 0.03 für M6G alleine auf 0.17 ± 0.08 für M6G mit PSC 833). Die antinozizeptiven Effekte von M6G in den Verhaltensexperimenten waren signifikant erhöht unter P-Glykoprotein Hemmung. Die Inhibition Probenecid sensitiver Transporter verursachte einen gleichmäßigen Anstieg der Blutplasma und Rückenmarkgewebekonzentration um den Faktor 1.4, ohne Veränderung des Konzentrationsverhältnis Rückenmark zu Blutplasma (0.081 ± 0.034 für M6G alleine, 0.082 ± 0.021 für M6G mit Probenecid). Die Verabreichung von Probenecid mit M6G zeigte eine signifikante Reduktion der Anzahl der Flinches im Formalinschmerztest um den Faktor 2.5 im Vergleich zu M6G alleine. Mit dieser Arbeit wurde gezeigt, dass Morphin-6-Glukuronid ein Substrat des Transporters P-Glykoprotein in Ratten ist. Die Verabreichung des P-Glykoprotein Inhibitors PSC 833 erhöhte die Penetration von M6G aus dem Blut in das zentrale Nervensystem und verstärkte dadurch seine antinozizeptive Wirkung. Dieser Effekt ist an der Blut-Hirn-Schranke lokalisiert. Weiterhin wurde demonstriert, dass der Blut-Hirn-Schranken Transport von Morphin-6-Glukuronid nicht durch Probenecid sensitive Transporter beeinflusst wird. Die Verabreichung von Probenecid verstärkte zwar die antinozizeptive Wirkung von M6G aufgrund eines Anstiegs der M6G Rückenmarkkonzentration, jedoch als Folge einer erhöhten Blutplasmakonzentration. Da M6G hauptsächlich in den Urin ausgeschieden wird, ist dieser Effekt wahrscheinlich in der Niere lokalisiert und vermindert die systemische Elimination von M6G. Somit ist M6G ein Substrat Probenecid sensitiver Transporter in Ratten, aber nicht an der Blut-Hirn-Schranke. Da viele Medikamente Inhibitoren von P-Glykoprotein oder Probenecid sensitiven Transportern sind, kann deren gleichzeitige Verabreichung mit Morphin bzw Morphin-6-Glukuronid zu erhöhten M6G ZNS Konzentrationen und Toxizität führen. Die Identifizierung dieser Transporter bedingten Interaktionen in unserer Arbeit kann helfen Medikamentenwechselwirkungen zwischen Inhibitoren und Morphin/ M6G zu vermeiden und die Patientensicherheit unter einer Morphin bzw. Morphin-6-Glukuronid Schmerztherapie zu steigern. Weitere Untersuchungen dieser Interaktionen im Menschen sind nötig, da sie klinisch von großer Bedeutung sind.
So wie der intermediale Dialog eine Polyphonie der Erzählerperspektiven zum Ausdruck bringt, die in 'Leben? Oder Theater?' narratologisches Programm ist, stellt die Serialität hier kein rein formales Experiment dar, sondern ist eng mit der Erzählung verbunden. Auch wenn es im Folgenden vorrangig um eine Serialität im Medium des Bildes geht, ist diese in eine Bilder'geschichte', mithin in einen narrativen Gesamtzusammenhang integriert und muss aus diesem und in Hinblick auf ihre Funktion für diesen untersucht werden. Es wird sich zeigen, dass Wiederholung und Serialität in 'Leben? Oder Theater?' thematisch immer an die Auseinandersetzung mit dem Tod bzw. das Spannungsverhältnis zwischen Leben und Tod gebunden ist.
Um Materie mit Nanometergenauigkeit anzuordnen, ist Selbstorganisation die mächtigste Strategie. DNA (Desoxyribonukleinsäure) ist hierfür ein hervorragendes Baumaterial, da sie ein billiges, programmierbares, biokompatibles und gut verstandenes Polymer ist. Aus diesen Gründen ist DNA zur Basis für ein schnell wachsendes Gebiet geworden: die DNA-Nanotechnologie. Quantification of circulating endothelial progenitor cells using the modified ISHAGE protocol

Aims: Circulating endothelial progenitor cells (EPC), involved in endothelial regeneration, neovascularisation, and determination of prognosis in cardiovascular disease can be characterised with functional assays or using immunofluorescence and flow cytometry. Combinations of markers, including CD34+KDR+ or CD133+KDR+, are used. This approach, however may not consider all characteristics of EPC. The lack of a standardised protocol with regards to reagents and gating strategies may account for the widespread inter-laboratory variations in quantification of EPC. We, therefore developed a novel protocol adapted from the standardised so-called ISHAGE protocol for enumeration of haematopoietic stem cells to enable comparison of clinical and laboratory data. Methods and Results: In 25 control subjects, 65 patients with coronary artery disease (CAD; 40 stable CAD, 25 acute coronary syndrome/acute myocardial infarction (ACS)), EPC were quantified using the following approach: Whole blood was incubated with CD45, KDR, and CD34. The ISHAGE sequential strategy was used, and finally, CD45dimCD34+ cells were quantified for KDR. A minimum of 100 CD34+ events were collected. For comparison, CD45+CD34+ and CD45-CD34+ were analysed simultaneously. The number of CD45dimCD34+KDR+ cells only were significantly higher in healthy controls compared to patients with CAD or ACS (p = 0.005 each, p<0.001 for trend). An inverse correlation of CD45dimCD34+KDR+ with disease activity (r = -0.475, p<0.001) was confirmed. Only CD45dimCD34+KDR+ correlated inversely with the number of diseased coronaries (r = -0.344; p<0.005). In a second study, a 4-week de-novo treatment of atorvastatin in stable CAD evoked an increase only of CD45dimCD34+KDR+ EPC (p<0.05). CD45+CD34+KDR+ and CD45-CD34+KDR+ were indifferent between the three groups. Conclusion: Our newly established protocol adopted from the standardised ISHAGE protocol achieved higher accuracy in EPC enumeration confirming previous findings with respect to the correlation of EPC with disease activity and the increase of EPC during statin therapy. The data of this study show the CD45dim fraction to harbour EPC.
The well-known proof of termination of reduction in simply typed calculi is adapted to a monomorphically typed lambda-calculus with case and constructors and recursive data types. The proof differs at several places from the standard proof. Perhaps it is useful and can be extended also to more complex calculi.
