"Goethe meint, dass es mein bestes Werk sei", schreibt Schiller an Körner am 13. Mai 1801. Bezeugt sind zudem die, vor allem von der studentischen Jugend, begeistert und bejubelt aufgenommenen ersten Aufführungen 1801 in Leipzig. Heute dürfte die Jungfrau eher zu den weniger oft aufgeführten Dramen Schillers gehören. Schließlich handelt es sich auf einen ersten unbefragten Blick um sein befremdlichstes, um ein bizarres und mysteriöses Stück. Warum beschäftigt sich der Aufklärer und Klassiker mit einer mythisch-mystischen Hexe und Heiligen (denn sie ist beides in der Geschichte) in einer romantischen Tragödie? Von den Romantikern sind wir allerhand Spuk- und Gespenstergeschichten gewöhnt und Zauberbäume und sprechende Heiligenbilder überraschen uns nicht. Aber Schiller? Schiller ist doch, so verstehe ich ihn jedenfalls, ein politischer Dichter. Und da liegt auch schon die Antwort: eben deshalb.
Der Impurismus ist eine literarische Strömung, die durch Jahrtausende geht, ein geistreiches, planvolles Spiel zur Produktion von hermetischer Literatur über ein tabuisiertes Thema (Sex). Zu dieser "littérature impure" gehört ein Geheimnis aus dem Urwissen der ältesten Kulturen, aus einer Zeit, in der Philosophie, Theologie und Kosmologie noch eins waren. Das alte Wissen wurde in den Bereichen konserviert, die bis heute von der konventionellen Wissenschaft als "esoterisch" ausgegrenzt werden (Astrologie, Kabbala, Tarot). Viele Autoren aber verschlüsseln ihre Texte mit der alten Lehre und verstecken sie hinter einer religiösen, spielerisch-humoristischen oder sozialkritisch engagierten Maske. Deshalb bleiben viele Texte trotz Interpretation hermetisch, besonders solche in der "Weltsprache der modernen Poesie" (Enzensberger). Eine neue Methode der literarischen Analyse (mit 57 Varianten der planvollen Verfremdung von Wörtern) kann die impuristische Literatur dekodieren. Dazu gehört als Raumordnung das alte "Weltbild der Windmühle". Dieses Literaturspiel wird als sublime Kulturtätigkeit aufgedeckt. Die Einzelseiten dieser Homepage können nur einige Einblicke in schwierige Zusammenhänge geben, die im langsamen Vortrag des Buches leichter zu verstehen sind. Auf beiden Wegen muß man sich Zeit nehmen und am besten einen philosophischen Wissenshunger mitbringen.
Background Translocations of the Mixed Lineage Leukemia (MLL) gene occur in a subset (5%) of acute myeloid leukemias (AML), and in mixed phenotype acute leukemias in infancy - a disease with extremely poor prognosis. Animal model systems show that MLL gain of function mutations may contribute to leukemogenesis. Wild-type (wt) MLL possesses histone methyltransferase activity and functions at the level of chromatin organization by affecting the expression of specific target genes. While numerous MLL fusion proteins exert a diverse array of functions, they ultimately serve to induce transcription of specific genes. Hence, acute lymphoblastic leukemias (ALL) with MLL mutations (MLLmu) exhibit characteristic gene expression profiles including high-level expression of HOXA cluster genes. Here, we aimed to relate MLL mutational status and tumor suppressor gene (TSG) methylation/expression in acute leukemia cell lines. Results Using MS-MLPA (methylation-specific multiplex ligation-dependent probe amplification assay), methylation of 24 different TSG was analyzed in 28 MLLmu and MLLwt acute leukemia cell lines. On average, 1.8/24 TSG were methylated in MLLmu AML cells, while 6.2/24 TSG were methylated in MLLwt AML cells. Hypomethylation and expression of the TSG BEX2, IGSF4 and TIMP3 turned out to be characteristic of MLLmu AML cell lines. MLLwt AML cell lines displayed hypermethylated TSG promoters resulting in transcriptional silencing. Demethylating agents and inhibitors of histone deacetylases restored expression of BEX2, IGSF4 and TIMP3, confirming epigenetic silencing of these genes in MLLwt cells. The positive correlation between MLL translocation, TSG hypomethylation and expression suggested that MLL fusion proteins were responsible for dysregulation of TSG expression in MLLmu cells. This concept was supported by our observation that Bex2 mRNA levels in MLL-ENL transgenic mouse cell lines required expression of the MLL fusion gene. Conclusion These results suggest that the conspicuous expression of the TSG BEX2, IGSF4 and TIMP3 in MLLmu AML cell lines is the consequence of altered epigenetic properties of MLL fusion proteins.
This study addresses the situation of the Sorbs, an indigenous minority living in Lusatia, Germany. Under the overarching rubric of ethnic and nationalist projects, women are usually symbolized as the guardians of culture and language. Women’s experiences as subjects in everyday life and in multi-facetted social practices therefore become veiled. The main concern in this book is to discuss how the women who identify themselves as/with Sorbs studied here construct their identities in the modern world, how they approach a sense of self and how they position themselves in their everyday lives, what kind of processes they undergo in their identity construction, and which factors are implicated in the formation of these identities. Investigating the Sorbian minority as the research subject and focusing on the female gender primarily involves intersections of ethnicity and gender, which are the points of departure for this study. As the research progresses, women’s gradual active construction of gender and ethnicity while living their everyday lives reveals a construction of multifarious and complex identifications across differences of gender, ethnicity, culture, religion and class. The results of research create Sorbian culture anew, craft Sorbian identity afresh and render the notion of Sorbian women in new terms. New meanings encased in these conceptions actually contain an active and transformative impetus. This thrust forces these ideas to undergo a process of redefinition. It is the life experiences people have in everyday practices that impel us to envisage identity construction as a dynamic, never-ending and open-ended articulation of one’s positionings.
The utilization of Ginkgo biloba in medicinal practice dates back to 1505 A.D. Ironically, the mechanisms of action of Ginkgo are not fully clarified till now. Nowadays, Ginkgo biloba leaf extracts are mainly indicated for mild to moderate cerebrovascular insufficiency and different forms of dementia. The fact that it is an herbal extract composed of several different components indeed adds to the intricacy of finding its mechanisms of actions. Indisputably, many scientists tried to elucidate the mechanisms of actions of Ginkgo. The first step to achieve this goal was to standardize the leaf extract. The standardized Ginkgo leaf extract contains 22-27 % flavonol glycosides, 2.8-3.4 % of ginkgolide A, B and C, as well as approximately 2.6-3.2 % bilobalide and below 5 ppm ginkgolic acids. A widespread standardized Ginkgo extract is the EGb 761, which was utilized in the current work. One of the earliest proposed mechanisms is the ability of the Ginkgo extract to act as an anti-oxidant, which could be explained by its high flavonoid contents. However, without doubt EGb 761 encompasses other characteristics which distinguish it from other herbal extracts that are also rich in flavonoids. Since free radicals and reactive oxygen species are highly associated with the mitochondrial functions, examination of the effect of EGb 761 on mitochondrial functions was lately addressed. Moreover, this was encouraged as the link between Alzheimer’s disease [AD] and the mitochondria started to emerge. Previously, our group observed mitochondrial protective actions of EGb 761 on cell culture in vitro. Furthermore, anti-apoptotic effects were previously described for EGb 761. However, only very few studies addressed the single constituents and their effect on mitochondrial functions. Flavonoids were studied in several other plant extracts and their radical scavenging activity is unquestionable, but EGb 761 has anti-apoptotic actions which may be attributed to its terpenoid fraction. Exclusively found in the Ginkgo plant, are the ginkgolides and therefore their actions are not yet fully elucidated. Moreover, those who attempted to address these constituents concentrated on one or two candidates, for example bilobalide or ginkgolide B and ignored the rest. Unfortunately, this led to incomplete results, and one couldn’t compare the relative activities of all EGb 761 components in order to state whether all the components are effective or not. ...
Algorithms and data structures constitute the theoretical foundations of computer science and are an integral part of any classical computer science curriculum. Due to their high level of abstraction, the understanding of algorithms is of crucial concern to the vast majority of novice students. To facilitate the understanding and teaching of algorithms, a new research field termed "algorithm visualisation" evolved in the early 1980's. This field is concerned with innovating techniques and concepts for the development of effective algorithm visualisations for teaching, study, and research purposes. Due to the large number of requirements that high-quality algorithm visualisations need to meet, developing and deploying effective algorithm visualisations from scratch is often deemed to be an arduous, time-consuming task, which necessitates high-level skills in didactics, design, programming and evaluation. A substantial part of this thesis is devoted to the problems and solutions related to the automation of three-dimensional visual simulation of algorithms. The scientific contribution of the research presented in this work lies in addressing three concerns: - Identifying and investigating the issues related to the full automation of visual simulations. - Developing an automation-based approach to minimising the effort required for creating effective visual simulations. - Designing and implementing a rich environment for the visualisation of arbitrary algorithms and data structures in 3D. The presented research in this thesis is of considerable interest to (1) researchers anxious to facilitate the development process of algorithm visualisations, (2) educators concerned with adopting algorithm visualisations as a teaching aid and (3) students interested in developing their own algorithm animations.
The goal of this research is to develop an understanding of what causes organizations and information systems to be “good” with regard to communication and coordination. This study (1) gives a theoretical explanation of how the processes of organizational adaptation work and (2) what is required for establishing and measuring the goodness of an organization with regard to communication and coordination. By leveraging concepts from cybernetics and philosophy of language, particularly the theoretical conceptualization of information systems as social systems and language communities, this research arrives at new insights. After discussing related work from systems theory, organization theory, cybernetics, and philosophy of language, a theoretical conceptualization of information systems as language communities is adopted. This provides the foundation for two exploratory field studies. Then a formal theory for explaining the adaptation of organizations via language and communication is presented. This includes measures for the goodness of organizations with regard to communication and coordination. Finally, propositions stemming from the theoretical model are tested using multiple case studies in six information system development projects in the financial services industry.
This thesis is devoted to the developement of a classical model for the study of the energetics and stability of carbon nanotubes. The motivation behind such a model stems from the fact that production of nanotubes in a well-controlled manner requires a detailed understanding of their energetics. In order to study this different theoretical approaches are possible, ranging from the computationally expensive quantum mechanical first principle methods to the relatively simple classical models. A wisely developed classical model has the advantage that it could be used for systems of any possible size while still producing reasonable results. The model developed in this thesis is based on the well-known liquid drop model without the volume term and hence we call it liquid surface model. Based on the assumption that the energy of a nanotube can be expressed in terms of its geometrical parameters like surface area, curvature and shape of the edge, liquid surface model is able to predict the binding energy of nanotubes of any chirality once the total energy and the chiral indices of it are known. The model is suggested for open end and capped nanotubes and it is shown that the energy of capped nanotubes is determined by five physical parameters, while for the open end nanotubes three parameters are sufficient. The parameters of the liquid surface model are determined from the calculations performed with the use of empirical Tersoff and Brenner potentials and the accuracy of the model is analysed. It is shown that the liquid surface model can predict the binding energy per atom for capped nanotubes with relative error below 0.3% from that calculated using Brenner potential, corresponding to the absolute energy difference being less than 0.01 eV. The influence of the catalytic nanoparticle on top of which a nanotube grows, on the nanotube energetics is also discussed. It is demonstrated that the presence of catalytic nanoparticle changes the binding energy per atom in such a way that if the interaction of a nanotube with the catalytic nanoparticle is weak then attachment of an additional atom to a nanotube is an energetically favourable process, while if the catalytic nanoparticle nanotube interaction is strong , it becomes energetically more favourable for the nanotube to collapse. The suggested model gives important insights in the energetics and stability of nanotubes of different chiralities and is an important step towards the understanding of nanotube growth process. Young modulus and curvature constant are calculated for single-wall carbon nanotubes from the paremeters of the liquid surface model and demonstrated that the obtained values are in agreement with the values reported earlier both theoretically and experimentally. The calculated Young modulus and the curvature constant were used to conclude about the accuracy of the Tersoff and Brenner potentials. Since the parameters of the liquid surface model are obtained from the Tersoff and Brenner potential calculations, the agreement of elastic properties derived from these parameters corresponds to the fact that both potentials are capable of describing the elastic properties of nanotubes. Finally, the thesis discuss the possible extension of the model to various systems of interest.
This thesis investigates the jet-medium interactions in a Quark-Gluon Plasma using a hydrodynamical model. Such a Quark-Gluon Plasma represents a very early stage of our universe and is assumed to be created in heavy-ion collisions. Its properties are subject of current research. Since the comparison of measured data to model calculations suggests that the Quark-Gluon Plasma behaves like a nearly perfect liquid, the medium created in a heavy-ion collision can be described applying hydrodynamical simulations. One of the crucial questions in this context is if highly energetic particles (so-called jets), which are produced at the beginning of the collision and traverse the formed medium, may lead to the creation of a Mach cone. Such a Mach cone is always expected to develop if a jet moves with a velocity larger than the speed of sound relative to the medium. In that case, the measured angular particle distributions are supposed to exhibit a characteristic structure allowing for direct conclusions about the Equation of State and in particular about the speed of sound of the medium. Several different scenarios of jet energy loss are examined (the exact form of which is not known from first principles) and different mechanisms of energy and momentum loss are analyzed, ranging from weak interactions (based on calculations from perturbative Quantum Chromodynamics, pQCD) to strong interactions (formulated using the Anti-de-Sitter/Conformal Field Theory Correspondence, AdS/CFT). Though they result in different angular particle correlations which could in principle allow to distinguish the underlying processes (if it becomes possible to analyze single-jet events), it is shown that the characteristic structure observed in experimental data can be obtained due to the different contributions of several possible jet trajectories through an expanding medium. Such a structure cannot directly be connected to the Equation of State. In this context, the impact of a strong flow created behind the jet is examined which is common to almost all jet deposition scenarios. Besides that, the transport equations for dissipative hydrodynamics are discussed which are fundamental for any numerical computation of viscous effects in a Quark-Gluon Plasma.
Das NANOG2-Gen ist ein Genduplikat des nur in embryonalen Stammzellen exprimierten Stammzellfaktors NANOG1, der eine Schlüsselrolle bei der Pluripotenz und der Selbsterneuerung der Stammzellen und möglicherweise der Krebsstammzellen hat. Zur Analyse, ob NANOG2 die beschriebenen Funktionen von NANOG1 in Gewebestammzellen und Krebsstammzellen übernimmt und ursächlich für die Leukämieentstehung verantwortlich ist, wurden embryonale Zellen und primäre Leukämiezellen auf NANOG2-Expression durch RT-PCR-Experimente, Western Blot Analysen und ChIP Assays untersucht. Dabei konnte eine Methode zur Unterscheidung der NANOG1 und NANOG2-Transkripte etabliert, neue Genstrukturen dieser Gene charakterisiert und NANOG2-Transkripte in hämatopoetischen Stammzellen und in allen primären Leukämiezellen detektiert werden. Außerdem konnte mit Hilfe von Genexpressionsanalysen eine äquivalente Funktion von NANOG2 zu NANOG1 festgestellt werden.
Asthma bronchiale ist eine obstruktive Lungenerkrankung, die mit einer Hyperreagibilität der Atemwege verbunden ist, ausgelöst durch eine Vielzahl von Stimuli. Es stellt die häufigste chronische Erkrankung im Kindesalter dar, ist jedoch bei Säuglingen und Kleinkindern nur schwer zu diagnostizieren. In diesem Alter stellen sehr häufig rezidivierende, teilweise durch (Virus-) Infekte ausgelöste obstruktive Bronchitiden die ersten Symptome eines Asthma bronchiale dar. In der vorliegenden Arbeit wurde an einem Patientenkollektiv von 172 Säuglingen und Kleinkindern (m:w = 115:57; Alter in Monaten = 13,1+/-11) mittels Bodyplethysmographie und Metacholin-Provokation untersucht, wie häufig einer rezidivierend obstruktiven Symptomatik eine bronchiale Hyperreagibilität (BHR) zugrunde liegt, und ob Kinder mit Risikofaktoren für ein Asthma bronchiale Unterschiede in der Lungenfunktion oder in der Reaktion auf eine Metacholin-Provokation zeigen im Vergleich zu Kindern ohne vorbestehende Risikofaktoren. Als Risikofaktoren galten Atopie, Allergie (erhöhte eosinophile Leukozyten, erhöhte IgE-Werte, positiver RAST) sowie eine Passivrauch-Exposition durch Tabakrauch der Eltern, die anhand eines Fragebogens ermittelt wurden. Insgesamt reagierten 26 der 108 Kinder mit RAST (24,1%) leicht- bis hochgradig, aber nur 14 der 108 Kinder (13,0%) mittel- bis hochgradig hyperreagibel, wobei ein Anstieg des Atemwegswiderstandes von mindestens 80% des Ausgangswertes, ein Abfall der Sauerstoffsättigung unter 95% sO2 oder eine klinische Symptomatik in Form von Husten und / oder Giemen, bzw. die Erfüllung mehrerer Kriterien als positive Reaktion gewertet wurde, gemäß den PD20-Leitlinien abhängig von der verabreichten Metacholin-Dosis. Hierbei zeigten Kinder mit atopischer Dermatitis tendenziell häufiger eine mittel- bis hochgradige BHR als das Vergleichskollektiv (mittel- bis hochgradige BHR: Kinder ohne Risikofaktoren= 14,1%; Kinder mit Atopie= 20,8%). Weitere Zusammenhänge zwischen Risikofaktoren für ein Asthma bronchiale und eine BHR konnten nicht festgestellt werden. Auch fand sich zwischen Tabakrauch-Exposition (exponierte Kinder: n=36; nicht exponierte Kinder: n=102) und der BHR keine Beziehung. Unsere Untersuchungen belegen, dass eine mittel- bis hochgradige Hyperreagibilität im Sinne eines Asthmasyndroms nur bei einem geringen Anteil (13,0%) der Kinder mit obstruktiven Bronchitiden nachweisbar ist. Entsprechend kritisch sollte der Einsatz von inhalativen Steroiden bei Kindern mit obstruktiven Zeichen gestellt werden.
In jüngster Zeit werden vom humanen Immundefizienzvirus-1-abgeleitete lentivirale Vektoren auch in der Gentherapie eingesetzt. Obwohl diese Vektoren nicht-mitotische Zellen transduzieren können, sind sie für einen Gentransfer in primäre ruhende Zellen oft nicht geeignet. In der Abteilung „Medizinische Biotechnologie“ des Paul-Ehrlich-Insituts wurde ein vom simianen Immundefizienzvirus SIVsmmPBj-abgeleiteter lentiviraler Vektor entwickelt, welcher im Gegensatz zu HIV-1-abgeleiteten Vektoren effizient in der G0-Phase des Zellzyklus arretierte humane Fibroblasten und humane primäre Monozyten transduzieren kann (Mühlebach et al., 2005). Im dieser Arbeit wurde das Potenzial dieses neuen Vektors für mögliche Anwendungen in der Gentherapie untersucht, indem seine Transduktionsfähigkeit für weitere primäre Zellen bestimmt wurde. Dabei waren humane hämatopoetische Stammzellen von besonderem Interesse, da sie die Vorläuferzellen aller Zellen des Blutes sind und die Eigenschaft zur Selbsterneuerung besitzen. Die Effizienz des Gentransfers in unstimulierten Stammzellen mit dem SIVsmmPBj-Vektor war jedoch nicht höher als mit anderen lentiviralen Vektoren. Interessanterweise konnte aber ein Einfluss der lentiviralen Vektoren auf das in vitro-Differenzierungspotenzial der transduzierten Stammzellen in die verschiedenen Vorläuferzellen beobachtet werden: Nach Transduktion mit dem SIVsmmPBj- und einem HIV-2-abgeleiteten Vektor differenzierten die Stammzellen bevorzugt in granulozytäre Vorläuferzellen, während die Transduktion mit einem HIV-1-abgeleiteten Vektor die Anzahl aller Vorläuferzellen deutlich reduzierte und insbesonders die Differenzierung in Makrophagenvorläuferzellen verminderte. Zur Untersuchung ihres Differenzierungs-potenzials in vivo wurden transduzierte hämatopoetische Stammzellen zur Repopulierung des Knochenmarks von NOD/SCID-Mäusen eingesetzt. Hierbei wurde jedoch kein Einfluss der verschiedenen lentiviralen Vektoren auf die Differenzierung der Stammzellen beobachtet. Allerdings konnte nur in einem sehr geringen Anteil der transplantierten Zellen eine Expression des übertragenen Gens nachgewiesen werden, so dass nicht ausgeschlossen werden kann, dass die transduzierten Zellen die Fähigkeit zur Repopulierung verloren hatten. Insgesamt ist jedoch zu sagen, dass entgegen der Erwartungen der neue Vektor keinen Vorteil gegenüber HIV-1-Vektoren zur Transduktion von hämatopoetischen Stammzellen aufweist. Weiter wurde die Transduktionsfähigkeit des SIVsmmPBj-Vekors für humanen B-Lymphozyten, Makrophagen und dendritische Zellen untersucht. Auf ruhenden B-Lymphozyten besaß der SIVsmmPBj-Vektor keinen Transduktionsvorteil gegenüber einem HIV-1-abgeleiteten Vektor, während Makrophagen und dendritische Zellen mit signifikant höherer Effizienz transduziert werden konnten. Die hohe Transduktionseffizienz des SIVsmmPBj-Vektors für Monozyten und dendritische Zellen eröffnet die Möglichkeit einer Anwendung in der Immuntherapie, da dendritische Zellen die professionellsten und effektivsten Antigen-präsentierenden Zellen sind. Daher wurde die generelle Eignung des SIVsmmPBj-Vektors für immuntherapeutische Anwendungen untersucht. Ein Tumor-assoziiertes Antigen (Mart-1) wurde in Monozyten übertragen und die transduzierten Zellen zu reifen dendritischen Zellen maturiert. Diese Zellen besaßen die Fähigkeit, Antigen-spezifische zytotoxische T-Zellen zu generieren, deren Funktion durch Sekretion von Zytokinen, in Einzelfällen auch durch spezifische Lyse von Mart-exprimierenden Tumorzellen nachgewiesen wurde. Weiterhin wurde gezeigt, dass nach Transduktion von Monozyten und deren Differenzierung zu Makrophagen auch diese prinzipiell in der Lage sind, Antigen-spezifische zytotoxische T-Zellen zu generieren. Obwohl hier keine vergleichenden Untersuchungen zur Effizienz des T-Zell-Primings durchgeführt werden konnten, ist die prinzipielle Eignung des SIVsmmPBj-abgeleiteten Vektors für eine Immuntherapie damit nachgewiesen. Schließlich wurde untersucht, ob die Maus oder nicht-menschliche Primaten als Tiermodelle für eine mögliche Weiterentwicklung des Vektors in Frage kommen. Murine Monozyten konnten jedoch nicht effizient transduziert werden. Hingegen erwies sich der SIVsmmPBj-Vektor als gut geeignet zur Transduktion von simianen Monozyten, so dass ein Affenmodell für Anwendungen des SIVsmmPBj-Vektors, wie beispielsweise zur Tumor-Immuntherapie oder für Vakzinierungsstudien, in Frage kommt.
Vasculogenesis as well as angiogenesis are important for postnatal development of blood vessels. Peripheral blood or bone marrow-derived endothelial precursor cells are used in clinical trials for therapeutic enhancement of postnatal neovascularization in patients suffering from coronary artery diseases. The vasculogenic potential of the precursor cell population depends on the appropriate retention of the infused cells to the ischemic tissue. However, cell-autonomous mechanisms regulating the attraction and retention of circulating cells in inflammatory tissue are not well understood. Caspases belong to a family of pro-apoptotic enzymes. Beyond cell death signals, caspase proteases additionally regulate non-apoptotic processes like cell morphology and migration in many cell types. The isoform Caspase-8 is essential for embryonal vasculogenesis in conditional knockout mice. In this study, we identified a novel apoptosis-unrelated role of Caspase-8 in circulating and bone marrow-derived cells for vascular repair. Caspase-8-specific inhibition abrogated the ex vivo formation of EPC from human peripheral blood. Moreover, Caspase-8 inhibition disables EPC migration and adhesion to different matrices and decreases the cell surface expression of the fibronectin receptor subunit integrin alpha 5 and the chemokine receptor CXCR4. In vitro and in vivo studies using bone marrow mononuclear cells derived from inducible Caspase-8- deficient mice revealed an essential role of Caspase-8 for EPC formation and neovascularization enhancing capacities of progenitor cells. Caspase-8 activity appears to be required for maintaining responses to matrix interaction and chemoattractants of EPC. Additional studies showed that the E3 ubiquitin ligase Cbl-b, a negative regulator of cell adhesion molecules including integrin alpha 5, is present in EPC at low protein levels under basal conditions, but markedly increases upon Caspase-8 inhibition. In vitro assays and overexpression studies in intact cells confirmed Caspase-8-dependent degradation of Cbl-b, providing a potential requirement for Caspase-8-regulated adhesion. Indeed, neovascularization of matrigel plugs was enhanced in mice lacking Cbl-b. Moreover, Cbl-b degradation in the presence of active Caspase-8 prevents the down-regulation of integrin alpha 5 and is associated with an enhanced vasculogenic activity of progenitor cells in hind limb ischemia. The identified upstream regulation of caspase-8 by cytokine IL-6 is only one possibility for fine-tuning the non-apoptotic enzymatic activity. In summary, this study shows a novel essential role of Caspase-8 for proper EPC adhesion-related signaling. Caspase-8 is involved in the function of adhesion molecules by regulation the E3 ubiquitin ligase Cbl-b. Strategies to improve survival of therapeutic injected progenitor cells by using caspase inhibitors should be addressed with caution. Because of the broad spectrum of activity of caspase-8, downstream targets of this caspase isoform and Cbl-b should be in more focus for therapeutic pretreatment to improve neovascularization of myocardial and ischemic tissue.
Word formation in Distributed Morphology (see Arad 2005, Marantz 2001, Embick 2008): 1. Language has atomic, non-decomposable, elements = roots. 2. Roots combine with the functional vocabulary and build larger elements. 3. Roots are category neutral. They are then categorized by combining with category defining functional heads.
This paper investigates the class of Tree-Tuple MCTAG with Shared Nodes, TT-MCTAG for short, an extension of Tree Adjoining Grammars that has been proposed for natural language processing, in particular for dealing with discontinuities and word order variation in languages such as German. It has been shown that the universal recognition problem for this formalism is NP-hard, but so far it was not known whether the class of languages generated by TT-MCTAG is included in PTIME. We provide a positive answer to this question, using a new characterization of TT-MCTAG.
Various concurrency primitives had been added to functional programming languages in different ways. In Haskell such a primitive is a MVar, joins are described in JoCaml and AliceML uses futures to provide a concurrent behaviour. Despite these concurrency libraries seem to behave well, their equivalence between each other has not been proven yet. An expressive formal system is needed. In their paper "On proving the equivalence of concurrency primitives", Jan Schwinghammer, David Sabel, Joachim Niehren, and Manfred Schmidt-Schauß define a universal calculus for concurrency primitives known as the typed lambda calculus with futures. There, equivalence of processes had been proved. An encoding of simple one-place buffers had been worked out. This bachelor’s thesis is about encoding more complex concurrency abstractions in the lambda calculus with futures and proving correctness of its operational semantics. Given the new abstractions, we will discuss program equivalence between them. Finally, we present a library written in Haskell that exposes futures and our concurrency abstractions as a proof of concept.
We present a CYK and an Earley-style algorithm for parsing Range Concatenation Grammar (RCG), using the deductive parsing framework. The characteristic property of the Earley parser is that we use a technique of range boundary constraint propagation to compute the yields of non-terminals as late as possible. Experiments show that, compared to previous approaches, the constraint propagation helps to considerably decrease the number of items in the chart.
Multicomponent Tree Adjoining Grammars (MCTAGs) are a formalism that has been shown to be useful for many natural language applications. The definition of non-local MCTAG however is problematic since it refers to the process of the derivation itself: a simultaneity constraint must be respected concerning the way the members of the elementary tree sets are added. Looking only at the result of a derivation (i.e., the derived tree and the derivation tree), this simultaneity is no longer visible and therefore cannot be checked. I.e., this way of characterizing MCTAG does not allow to abstract away from the concrete order of derivation. In this paper, we propose an alternative definition of MCTAG that characterizes the trees in the tree language of an MCTAG via the properties of the derivation trees (in the underlying TAG) the MCTAG licences. We provide similar characterizations for various types of MCTAG. These characterizations give a better understanding of the formalisms, they allow a more systematic comparison of different types of MCTAG, and, furthermore, they can be exploited for parsing.
Background Chemoresistance acquisition may influence cancer cell biology. Here, bioinformatics analysis of gene expression data was used to identify chemoresistance-associated changes in neuroblastoma biology. Results Bioinformatics analysis of gene expression data revealed that expression of angiogenesis-associated genes significantly differs between chemosensitive and chemoresistant neuroblastoma cells. A subsequent systematic analysis of a panel of 14 chemosensitive and chemoresistant neuroblastoma cell lines in vitro and in animal experiments indicated a consistent shift to a more pro-angiogenic phenotype in chemoresistant neuroblastoma cells. The molecular mechanims underlying increased pro-angiogenic activity of neuroblastoma cells are individual and differ between the investigated chemoresistant cell lines. Treatment of animals carrying doxorubicin-resistant neuroblastoma xenografts with doxorubicin, a cytotoxic drug known to exert anti-angiogenic activity, resulted in decreased tumour vessel formation and growth indicating chemoresistance-associated enhanced pro-angiogenic activity to be relevant for tumour progression and to represent a potential therapeutic target. Conclusions A bioinformatics approach allowed to identify a relevant chemoresistance-associated shift in neuroblastoma cell biology. The chemoresistance-associated enhanced pro-angiogenic activity observed in neuroblastoma cells is relevant for tumour progression and represents a potential therapeutic target.
Die vorliegende Arbeit untersucht ausgewählte Eigenschaften von Preferential Attachment-Graphen. Darunter verstehen wir eine Klasse komplexer zufälliger Graphen, die mit einer vorgegebenen Konfiguration gestartet werden und anschließend mit jedem Zeitschritt um eine Ecke und m Kanten wachsen. Die Wachstumsregeln sind so gestaltet, dass eine neue Ecke ihre Kanten bevorzugt an Ecken sendet, die bereits mit vielen anderen Ecken verbunden sind, woraus sich die Bezeichnung Preferential Attachment (PA) ableitet. Die Arbeit stellt zunächst heuristisch die Eigenschaft der Skalenfreiheit von PA-Modellen vor und bespricht anschließend einen Beweis zu dieser These. Weiter betrachten wir den Durchmesser von PA-Graphen und untersuchen das Verhalten bei Anwachsen des Graphen. Wir erkennen, dass der Durchmesser bei wachsendem Graphen deutlich langsamer wächst, was wir als Small World-Phänomen bezeichnen. Die zentralen Aussagen und Beweise orientieren sich an den Arbeiten von Remco van der Hofstad, der die bekannten PA-Modelle um einen Parameter erweitert hat. Damit ist es möglich, sowohl logarithmische als auch doppelt-logarithmische Schranken für den Durchmesser zu erhalten.
Die Fluoreszenz der organischen Verbindung ''Pigment Yellow 101'' wurde mittels zeitaufgelöster Anreg-Abtast-Spektroskopie im sichtbaren Spektralbereich untersucht. P.Y. 101 und ein Derivat mit zusätzlichen Methylgruppen an den Azin-Kohlenstoffatomen, wurden mit zwei Derivaten verglichen, die keine Hydroxygruppen tragen und nicht fluoreszieren. Es konnte gezeigt werden, dass die Fluoreszenz bei diesen Verbindungen eine intrinsische Eigenschaft ist, die von der Reihenfolge der angeregten Zustände bestimmt wird. Bei Derivaten mit Hydroxygruppe entspricht der niedrigste angeregte Zustand einem pp*-Übergang, welcher hauptsächlich durch Fluoreszenz zurück in den Grundzustand gelangt. Bei den Verbindungen ohne Hydroxygruppe fehlt die stabilisierende Wasserstoffbrücke, die zur Absenkung des entsprechenden Zustands führt, so dass bei diesen Derivaten der unterste angeregte Zustand einem np*-Übergang entspricht, der optisch verboten ist und somit nicht direkt populiert wird. Bei der Anregung wird ein höher angeregter Zustand bevölkert, der in den niedrigsten angeregten Zustand relaxiert, welcher über eine konische Durchschneidung schnell und strahlungsfrei erreicht werden kann. Der niedrigste angeregte Zustand schneidet seinerseits den Grundzustand, so dass auch die Repopulierung des Ausgangszustands schnell, effizient und strahlungslos abläuft. Eine ganz andere Art der Photochemie wird bei Azobenzol (AB) und AB-Derivaten beobachtet. Durch Isomerisierung um die N-N-Doppelbindung wird ein Photoprodukt gebildet, das cis-Isomer. Die Quantenausbeute der Isomerisierung ist dabei abhängig davon, welcher elektronische Übergang angeregt wurde. Substituenten am AB üben unterschiedlichen Einfluss auf die elektronischen Eigenschaften aus: Elektronegative Gruppen in meta-Position zur Diazobrücke verändern die Lage der Absorptionsbanden kaum, ebensowenig die Rate der thermischen Rückisomerisierung und auch die Dynamik nach Photoanregung zeigt keine signifikanten Unterschiede in Abhängigkeit vom Substituenten und kann in Analogie an unsubstituiertes AB interpretiert werden. Nach Anregung des np*-Übergangs zerfällt der angeregte Zustand biexponentiell. Die Anregung des pp*-Übergangs, dem optisch erlaubten Übergang, führt zur Population des zweiten angeregten Zustands. Entlang einer konischen Durchschneidung wird schnell und effizient der erste angeregte Zustand populiert. Dieser Prozess zeigt sich bei den zeitaufgelösten Messungen in einer schnellen Zerfallszeit, die nur wenige hundert Femtosekunden beträgt. Die nun folgenden Prozesse finden im ersten angeregten Zustand statt und sind vergleichbar mit den Prozessen nach np*-Anregung. Der Hauptunterschied zwischen den beiden Dynamiken im S1-Zustand ist die Ausgangsgeometrie, mit der die Moleküle diesen Zustand populieren (Franck-Condon-Bereich bei direkter Anregung, bzw. eine große Bandbreite an Geometrien nach dem S2-S1-Übergang), sowie die Schwingungsenergie die das System jeweils besitzt. Auch nach ursprünglicher S2-Anregung können zwei Zerfallszeiten dem ersten angeregten Zustand zugeordnet werden, die analog der direkten Population als Geometrieänderung in Richtung des Potentialminimums und Isomerisierung bzw. Rückkehr in den Grundzustand interpretiert werden und in der Größenordnung von einer bzw. fünf Pikosekunden liegen. Die Modifizierung von Peptiden mit Azobenzol stellt einen Weg dar, zeitlich synchronisiert, eine Störung in der Struktur zu induzieren. Dieses Konzept wurde bereits erfolgreich zur Untersuchung der Faltung zyklischer Peptide eingesetzt. Modifizierte Kollagenstränge sind eine Erweiterung dieses Prinzips, da sie eine Möglichkeit darstellen, die Tertiärstruktur eines Peptids zu adressieren. Für das vorliegende Azokollagen konnte gezeigt werden, dass die gewählte Aminosäuresequenz auch nach Anbringen der Azobenzolklammer zu gefaltetem Kollagen führt, welches thermodynamisch stabil ist. Der Schmelzpunkt der Tripelhelix liegt unter den gewählten Bedingungen bei 55°C. Die Isomerisierung des Azobenzols induziert eine Störung der Tertiärstruktur, die zur teilweisen Entfaltung der Tripelhelix führt, was ein reversibler Prozess ist. Eine vollständige Entfaltung findet erst bei Temperaturen oberhalb des Schmelzpunkts statt. Zeitaufgelöste Messungen im sichtbaren Spektralbereich zeigten, dass die Schalterdynamik durch Anbringung an das Peptid nicht signifikant im Vergleich zum reinen AB-Schalter verändert wird.
Ausgangspunkt dieser Dissertation ist die Frage: „Was ist fundamentaler als die Raum-Zeit?“ Nach Immanuel Kant sind aber Raum & Zeit als reine Formen der sinnlichen Anschauung „a priori“ aller Erfahrung und allen Denkens und konstituieren somit die „Bedingungen, unter denen Erfahrung erst möglich ist“. Im Jahre 1941 hat Konrad Lorenz diese Apriori biologisch interpretiert und damit das Konzept einer Evolutionären Erkenntnislehre formuliert. Deren Hauptthese lautet: „Unser Erkenntnis-Apparat ist ein Ergebnis der Evolution. Die Subjektiven Erkenntnisstrukturen passen auf die Welt, weil sie sich im Laufe der Evolution in Anpassung an eben diese reale Welt herausgebildet haben. Sie stimmen mit den realen Strukturen (teilweise) überein, weil nur eine solche Übereinstimmung das Überleben ermöglichte.“ Wendet man dies auf die Erfahrung von Raum & Zeit an, so gelangt man zur Kognitionswissenschaft von Raum & Zeit. Das Ergebnis der Analyse der Kognition von Raum & Zeit lässt sich wie folgt zusammenfassen. Im Kognitiven System des Menschen gibt es mehrere Informationsrepräsentationssysteme. Ein Sprachlichlogisches Repräsentationssystem, ein Nonverbales Repräsentationssystem zur Speicherung von Multimodalen Spatiotemporalen Informationen und ein Internes Repräsentationssystem zur Speicherung von Subjektiven Informationen über interne Körperzustände. Das Multimodal-Spatiotemporale Repräsentationssystem lässt sich dann weiter in Modalitätsspezifische Repräsentationssysteme für die einzelnen Sinnesmodalitäten und ein Amodales Repräsentationssystem für Spatiotemporale Information untergliedern. Letzteres ist weiter in Amodale Repräsentationssysteme für Reinräumliche Informationen, Spatiotemporale Informationen & Reinzeitliche Informationen untergliedert. Mit dem Reintemporalen Repräsentationssystem hat dieses Amodale Repräsentationssystem auch Anteil am Internen Körperrepräsentationssystem. Für die Kognition der Zahlen kommt nur noch ein Zahlenrepräsentationssystem für die Zahlen 1-3 als eigenständiges Repräsentationssystem hinzu. Alle anderen Zahlenrepräsentationen benutzen das Räumliche oder das Sprachlichlogische Repräsentationssystem. Alle diese Repräsentationssysteme sind das Ergebnis von Phylogenetischen, Ontogenetische & Psychogenetischen Entwicklungsprozessen. Aus diesen Erkenntnissen wird anschließend untersucht, naturphilosophische Konsequenzen für die Raum-Zeit-Physik ergeben. Insbesondere geht es um die Frage von Gerhard Vollmer, die lautet: „Nach der Evolutionären Erkenntnistheorie sind die subjektiven Strukturen des Erkenntnis-Apparates für die Erkenntnis konstitutiv. Gilt dies für alle Stufen des Erkenntnis-Prozesses?“ Das Ergebnis dieser Analyse ist, dass das Visuospatiotemporale Informationsrepräsentationssystem die Kognitive Basis der Physikalischen Theorien mit Klassischem Materiekonzept bildet, mit dessen Hilfe das Visuell-Nonverbale Sensomotorische System diese Theorien mit Hilfe von Kognitionsprozessen aus Interaktionen mit der Physikalischen Außenwelt konstruiert hat. Darüber hinaus beruhen diese Theorien auch auf dem Sprachlichlogischen Informationsrepräsentations-System als Kognitiver Basis und sind das Produkt von Kognitiven Math. Basisfähigkeiten, mit deren Hilfe sie ebenfalls aus Interaktionen mit der Physikalischen Außenwelt abgeleitet wurden. Dabei dominiert das Visuospatiotemporale Informationsrepräsentationssystem in der Kognitiven Basis gegenüber dem Sprachlichlogischen Informationsrepräsentationssystem eindeutig! Auch die Quantenmechanik hat diese beiden Informationsrepräsentationssysteme als Kognitive Basis. Allerdings dominiert hier das Sprachlichlogische Informationsrepräsentationssystem gegenüber dem Visuospatiotemporalen Informationsrepräsentationssystem! Da das Sprachlichlogische Informationsrepräsentationssystem aber ein Symbolisches Informationsrepräsentationssystem ist, bedarf es zu seiner Funktionsfähigkeit der Abbildung seiner Symbole auf Informationsstrukturen des Multimodalen Spatiotemporalen Informationsrepräsentationssystems, damit die mit seiner Hilfe repräsentierten Sprachlichlogischen Informationen überhaupt Teil des Informationellen Modells der Außenwelt des Kognitiven Systems sein können. Damit lässt sich die oben gestellte Frage wie folgt beantworten: Ja, diese Kognitiven Strukturen sind nicht nur für die Ebenen der Wahrnehmungs- & Erfahrungserkenntnis konstitutiv, sondern für alle Stufen des Erkenntnisprozesses. Das heißt: „Auch die Wissenschaftliche Erkenntnis ist biologisch/genetisch determiniert!“
The study of the electromagnetic structure of hadrons plays an important role in understanding the nature of matter. In particular the emission of lepton pairs out of the hot and dense collision zone in heavy-ion reactions is a promising probe to investigate in-medium properties of hadrons and in general the properties of matter under such extreme conditions. The first experimental observation of an enhanced di-electron yield in the invariant-mass region 0:3 - 0:7 GeV/c2 in p+Be collisions at 4:9 GeV/u beam energy [2] was announced by the DLS collaboration [1]. Recent results of the HADES collaboration show a moderate enhancement above n Dalitz decay contributions for 12C+12C at 1 and 2 GeV/u [3, 4] confirming the DLS results. There are several theoretical explanations of this observation, most of them focusing on possible in-medium modifications of the properties of vector mesons. At low beam energies the question whether the observed excess is related to any in-medium effects remains open because of uncertainties in the description of elementary di-electron sources. In this work the di-electron production in p+p and d+p reactions at a kinetic beam energy of 1:25 GeV/u measured by the HADES spectrometer is discussed. At Ekin = 1:25 GeV/u, i.e. below the n meson production threshold in proton-proton reactions, the delta Dalitz decay is expected to be the most abundant source above the pi 0 Dalitz decay region. The observed large difference in di-electron production in p+p and d+p collisions suggests that di-electron production in the d+p system is dominated by the n+p interaction. In order to separate delta Dalitz decays and np bremsstrahlung the di-electron yield observed in p+p and n+p reactions, both measured at the same beam energy, has been compared. The main interest here is the investigation of iso-spin effects in baryonic resonance excitations and the off-shell production of vector mesons [5]. We indeed observe a large difference in di-electron production in p+p and n+p reactions. Results of these studies will be compared to recent calculations. We will also present our experimentally defined cocktail for heavy-ion data. At much higher beam energies experimental results of the CERES [6] and NA60 [7] collaborations also show an enhancement in the invariant mass region 0:3 - 0:7 GeV/c2, in principle similar to the situation in DLS. A strong excess of lepton pairs observed by recent high energy heavy-ion dilepton experiments hint to a strong influence of baryons, however no data exist at highly compressed baryonic matter, achievable in heavy-ion collisions from 8 - 45 GeV/u beam energy. These conditions would allow to study the expected restoration of chiral symmetry by measuring in-medium modifications of hadronic properties, an experimental program which is foreseen by the future CBM experiment at FAIR. The experimental challenge is to suppress the large physical background on the one hand and to provide a clean identification of electrons on the other hand. In this work, strategies to reduce the combinatorial background in electron pair measurements with the CBM detector are discussed. The main goal is to study the feasibility of effectively reducing combinatorial background with the currently foreseen experimental setup, which does not provide electron identification in front of the magnetic field.
This paper gives a brief overview of computation models for data stream processing, and it introduces a new model for multi-pass processing of multiple streams, the so-called mp2s-automata. Two algorithms for solving the set disjointness problem with these automata are presented. The main technical contribution of this paper is the proof of a lower bound on the size of memory and the number of heads that are required for solving the set disjointness problem with mp2s-automata.
Ich werde allerdings noch zur Diskussionskultur in den USA gegen Ende meines Vortrages zu sprechen kommen. Heute möchte ich über meine aktuelle Studie zum Münzhandel in Nordamerika referieren. Obwohl ich mein Hauptaugenmerk auf die Situation in den USA lenke, sind selbstverständlich viele Aspekte auf Europa übertragbar. Zuerst möchte ich den Handel mit Neufunden darlegen. Es ist wichtig darauf hinzuweisen, dass sich viele Händler und Sammler, besonders in den Vereinigten Staaten, darauf berufen, dass der Handel mit antiken Münzen in keinem Zusammenhang mit der Raubgräberei und dem Handel anderer antiker Objekte steht. Danach möchte ich den Umfang des Handels mit Neufunden besprechen und aufzeigen, wie Münzen aus Raubgrabungen über illegale Transaktionen und kriminelle Strukturen von der Ausgrabungsstätte über Schmuggler, Großhändler und spezialisierte Händler schließlich zu den Sammlern gelangen. Ich hoffe, wir stimmen alle dahingehend überein, dass die Probleme eines nicht-regulierten Handels sowohl für die Wissenschaft als auch für die Sammler verheerend sein können. Daher ist es im Interesse beider Seiten diesbezüglich zu kooperieren. Ich bin sehr zuversichtlich, dass das Verhältnis von Sammlern, Händlern und Archäologen in Deutschland ebenso wie im gesamten Europa größere Möglichkeiten für einen ehrlichen Dialoge und positive Änderungen zulassen als in den USA, worauf ich ebenfalls eingehen möchte. ...
TNFAIP3 (A20) is a tumor suppressor gene in Hodgkin lymphoma and primary mediastinal B cell lymphoma

Proliferation and survival of Hodgkin and Reed/Sternberg (HRS) cells, the malignant cells of classical Hodgkin lymphoma (cHL), are dependent on constitutive activation of nuclear factor {kappa}B (NF-{kappa}B). NF-{kappa}B activation through various stimuli is negatively regulated by the zinc finger protein A20. To determine whether A20 contributes to the pathogenesis of cHL, we sequenced TNFAIP3, encoding A20, in HL cell lines and laser-microdissected HRS cells from cHL biopsies. We detected somatic mutations in 16 out of 36 cHLs (44%), including missense mutations in 2 out of 16 Epstein-Barr virus–positive (EBV+) cHLs and a missense mutation, nonsense mutations, and frameshift-causing insertions or deletions in 14 out of 20 EBV– cHLs. In most mutated cases, both TNFAIP3 alleles were inactivated, including frequent chromosomal deletions of TNFAIP3. Reconstitution of wild-type TNFAIP3 in A20-deficient cHL cell lines revealed a significant decrease in transcripts of selected NF-{kappa}B target genes and caused cytotoxicity. Extending the mutation analysis to primary mediastinal B cell lymphoma (PMBL), another lymphoma with constitutive NF-{kappa}B activity, revealed destructive mutations in 5 out of 14 PMBLs (36%). This report identifies TNFAIP3 (A20), a key regulator of NF-{kappa}B activity, as a novel tumor suppressor gene in cHL and PMBL. The significantly higher frequency of TNFAIP3 mutations in EBV– than EBV+ cHL suggests complementing functions of TNFAIP3 inactivation and EBV infection in cHL pathogenesis.
In the New-Keynesian model, optimal interest rate policy under uncertainty is formulated without reference to monetary aggregates as long as certain standard assumptions on the distributions of unobservables are satisfied. The model has been criticized for failing to explain common trends in money growth and inflation, and that therefore money should be used as a cross-check in policy formulation (see Lucas (2007)). We show that the New-Keynesian model can explain such trends if one allows for the possibility of persistent central bank misperceptions. Such misperceptions motivate the search for policies that include additional robustness checks. In earlier work, we proposed an interest rate rule that is near-optimal in normal times but includes a cross-check with monetary information. In case of unusual monetary trends, interest rates are adjusted. In this paper, we show in detail how to derive the appropriate magnitude of the interest rate adjustment following a significant cross-check with monetary information, when the New-Keynesian model is the central bank’s preferred model. The cross-check is shown to be effective in offsetting persistent deviations of inflation due to central bank misperceptions. Der Gesetzgeber hat bei der Entstehung des SoFFin auf bekannte Gesetzesmodelle zurück gegriffen. Sondervermögen sind grundsätzlich keine juristischen Personen und auch nicht rechtsfähig. Der SoFFin ist gleichwohl rechtlich selbstständig und damit im Sinne des öffentlichen Rechts „teilrechtsfähig“. Er hat keine eigenen Organe, vielmehr wird er von der FMSA verwaltet und vertreten. Die FMSA wurde als rechtliche unselbständige Anstalt des öffentlichen Rechts bei der Deutschen Bundesbank errichtet. Das daraus entstandene organisatorisch kaum einzuordnende Konstrukt lässt viele Fragen offen. Es verstößt nicht gegen Verfassungsrecht, solange die Unabhängigkeit der Bundesbank gewahrt bleibt. Wegen ihrer neu übernommenen Aufgabe aus § 8a FMStFG (Errichten von bundesrechtlichen Abwicklungsanstalten) wurde die FMSA nunmehr, im Juli 2009, in eine bundesunmittelbare, rechtsfähige Anstalt des öffentlichen Rechts im Geschäftsbereich des Bundesministeriums der Finanzen umgewandelt. Sie ist ein eigener Verwaltungsträger und kann bei der Abwicklung ihrer Geschäfte die Deutsche Bundesbank in Anspruch nehmen. Die FMSA unterliegt der Rechts- und Fachaufsicht des Bundesfinanzministeriums. Das Organ der FMSA ist der Leitungsausschuss. Als Verwaltungsorgan hat er die Anstalt nach außen und vor Gericht zu vertreten. Eine genaue Zuweisung weiterer Kompetenzen und Aufgaben ist im Gesetz nicht vorgesehen. Zumindest ist er für alle Entscheidungen zuständig, die nicht dem Bundesministerium der Finanzen oder dem Lenkungsausschuss zugewiesen sind. Der Lenkungsausschuss ist ein von der FMSA unabhängiger, interministerieller Ausschuss. Er ist zum einen eine der Kontrollinstanzen für die FMSA und zum anderen kommt ihm Entscheidungsbefugnis im Aufgabenbereich der FMSA zu. Wie das Verhältnis des Lenkungsausschusses zum Bundesministerium der Finanzen zu sehen ist, ist unklar. Die parlamentarische Aufsicht über Maßnahmen, die vor allem Schulden des Fonds betreffen, wird darüber hinaus vom „Gremium zum Finanzmarktstabilisierungsfonds“ sichergestellt. Haushaltsrechtlich wirft der Fonds einige Fragen auf. Allerdings ist seine Errichtung in Form eines Sondervermögens in der Gesamtbetrachtung verfassungsrechtlich nicht bedenklich. Auch unterliegt der SoFFin einer umfassenden Prüfung durch den Bundesrechnungshof. Im Hinblick auf die Vergabe von Stabilisierungshilfen an antragstellende Unternehmen kann die FMSA in zweierlei Formen handeln: Zum einen öffentlich-rechtlich und zum anderen privatrechtlich. Die FMSA kann sich allerdings nur ausnahmsweise privatrechtlicher Handlungsformen bedienen. Auch in diesem Fall darf sie nicht willkürlich handeln, sondern ist an die Grundsätze des Verwaltungsprivatrechts gebunden.
Azopeptide: Peptide mit eingebauten lichtgesteuerten Schaltern sind interessante Systeme, um konformationelle Dynamik in Peptiden zu untersuchen. In dieser Arbeit ist es gelungen einen solchen Schalter herzustellen und in ein von Robertson et al. entworfenes Modellsystem als Teil des Peptidrückgrats einzuführen. Es wurde somit die Synthese von Peptiden mit eingebauten lichtgesteuerten Schaltern fortgeführt und auf ein größeres System übertragen. Die zu erwartenden Probleme bei der Synthese eines Systems dieser Größe (30 Aminosäuren + Schalter) konnten durch Modifizierung der Standardsynthese für Peptide (Fmoc-Strategie) an der Festphase erreicht werden. Es war daher möglich, ausreichende Mengen des Peptids herzustellen sowie die freie SH-Gruppe des Peptids mit einer Schutzgruppe zu versehen, was dem Molekül zu weiterer Stabilität verhalf. Das Azopeptid wurde mit UV/vis- und Ultrakurzzeit-Spektroskopie, und besonders im Vergleich mit dem Schalter AMPB alleine, charakterisiert. Hierbei wurden folgende Erkenntnisse offen gelegt: - Das Azopeptid in Wasser verhält sich bei Belichtung (367 nm) sehr ähnlich dem AMPB (7) in DMSO (isosbestischer Punkt bei 288 nm) - Die thermische Rückreaktion lässt sich bei 330 nm biexponentiell fitten, bei 260 nm nicht, was Rückschlüsse auf mangelnde Stabilität des Azopeptids nach Belichtung zulässt (freie SH-Gruppe). - Der Abfall des angeregten Zustandes des Azopeptids folgt multiexponentiellen Kinetiken auf Zeitskalen zwischen einigen hundert fs bis zu wenigen ps. - Der Schalter AMPB (7) in DMSO verhält sich bei Belichtung (367 nm) sehr ähnlich dem beidseitig entschütztem Schalter (8) in Wasser. - Es sehr ähnliche Kinetiken für AMPB (7) in DMSO und das Azopeptid in Wasser über den gesamten spektralen Bereich werden gefunden; Absorptionsaufbau erfolgt innerhalb der Zeitauflösung des Experiments, Unterschied um einen Faktor 2 in der Zerfallsdynamik, die für das Azopeptid langsamer ist. Parvulustat: Parvulustat ist wie Tendamistat ein alpha-Amylase-Inhibitor; die Struktur von Tendamistat ist bereits sehr gut sowohl durch NMR als auch durch Röntgenkristallographie untersucht ist. Mit Parvulustat teilt Tendamistat nur 29,6 % Sequenzidentität bei ähnlicher Länge und gleicher Funktion der beiden Proteine. Es war daher von großem Interesse, die Struktur von Parvulustat aufzuklären um Ähnlichkeiten und Unterschiede der beiden Proteine diskutieren zu können. In dieser Arbeit ist es gelungen mit Hilfe der hochauflösenden, heteronuklearen 3D NMR-Spektroskopie in Lösung und iterativen Rechungsmethoden die Struktur des Proteins Parvulustat, anhand von 15N- und 13C,15N-markierten Proben, in sehr guter Qualität aufzuklären. Weiterhin ist es gelungen, dynamische Eigenschaften des Proteins durch Relaxationsdaten darzustellen. Basierend auf diesen Daten war es möglich die beiden Proteine Parvulustat und Tendamistat umfassend miteinander zu vergleichen und Schlüsse bezüglich ihres Bindungsmechanismus zu ziehen. Insgesamt ist zwischen beiden Proteinen eine große Ähnlichkeit zu verzeichnen, aber es wurden auch einige Unterschiede festgestellt: beide Proteine besitzen zwar die gleiche beta-Faltblatt-Struktur, jedoch sind bei Parvulustat die einzelnen Stränge etwas kürzer ausgebildet. Weiterhin hat in Parvulustat ein Strang eine andere Krümmung, weil ein Prolin anstelle eines Leucins in Tendamistat sitzt und durch seine einzigartige Form die Struktur in dieser Region ändert. Bezug nehmend auf die Ladungsverteilung beider Proteine ist festzustellen, dass beide durch ein hydrophobes Herzstück stabilisiert werden und sich insgesamt sehr ähnlich sind, bis auf die Position R44 in Parvulustat bzw. Y46 in Tendamistat, was in Parvulustat eine positive Ladung an der Oberfläche generiert. Generell ist noch zu sagen, dass man beim Interpretieren der Daten in Bezug auf Tendamistat vorsichtig sein muss, da es durchaus Unterschiede in der Datenakquise gibt: im Gegensatz zu Tendamistat dessen Struktur anhand von homonuklearen 2D NMR-Techniken aufgeklärt wurde, waren für Parvulustat bereits 3D Pulssequenzen verfügbar, NMR-Spektrometer mit höheren Feldern, weiterentwickelte Rechenprogramme, so dass u. a. auch die Relaxationsdaten einflussnehmend in die Strukturrechnung mit eingebaut werden konnten.
Despite the well-known importance of ribonucleic acids (RNA) in cell biology, it is astounding to realize the pace at which new fundamental functions of RNAs have been discovered. One of the fundamental reasons for the multitude of functions of RNA is the property of RNA to adopt different conformations or folds. The primary sequence of RNA, a linear polymer built from four different repetition units, can fold into alternate secondary structure motifs which in turn form alternate long-range interactions in complex tertiary structures. Ligands such as metal ions or small molecular weight metabolites and also proteins or peptides can bind to RNA and induce the changes in tertiary conformation. For example, in the cell, RNA participates in gene regulation in the form of riboswitches. Riboswitches are found in untranslated regions of messenger RNA (mRNA) and adopt alternate conformations depending on the presence or absence of specific metabolites. If a metabolite is present above a specific concentration, it induces a conformational change in the respective riboswitch by binding and thereby alters gene expression. Another example is the RNA thermometer which participates in the cell translational mechanism by a similar strategy. Translation initiation requires the binding of RNA thermometers to the ribosome. The ribosome binding region is located in the 5’ untranslated region of mRNA. At low temperatures this region is prevented from binding to the ribosome by forming basepairs. At higher temperatures, these basepairs dissociate allowing ribosome binding and subsequent translation. Therefore, the characterization and delineation of the kinetics and pathway of RNA folding is important to understand the function of RNA and is an important contribution to fundamentally understand RNA’s role in the cell. RNA conformational transitions occur over a wide range of timescales. Depending on the timescale, various biophysical techniques are used to study RNA conformational transitions. In these biophysical studies, achieving good structural and temporal resolution constitute frequently encountered challenges or limitations. For example, single molecule FRET spectroscopy provides high temporal resolution in the milliseconds at high sensitivity but lacks atomic resolution. Recent advances in the field of Nuclear Magnetic Resonance (NMR) spectroscopy have enabled the elucidation of tertiary folding events to be characterized with atomic resolution. This thesis involves the use of NMR spectroscopy to characterize the folding of RNA molecules. Kinetics experiments require rapid initiation of the kinetics followed by monitoring of the reaction. In this thesis, two different folding initiation techniques have been applied and coupled to the subsequent detection of RNA folding using NMR spectroscopy, namely, photocaging and rapid mixing. The method of photocaging is well established (Kuhn and Schwalbe, 2000) and builds on the following principle: A photolabile moiety is attached to a molecule that prevents a specific interaction. Upon irradiation of the molecule with the photolabile group using laser light at a specific wave length, at which the molecule of interest is not absorbing, the protecting group is released. In our group, together with the group of S. Pitsch, ETH Lausanne, we could "cage" RNA at its equilibrium state by a photolabile molecule (similar work has been carried out in the group of A. Heckel). Rapid and traceless release of the photolabile precursor compound by a laser pulse releases the RNA to fold into its native state; the build-up of the native state of the RNA is monitored by NMR signals that are uniquely characteristic for the native state of the RNA. By optically coupling a laser source to an NMR magnet, the above procedure can take place in situ and the kinetics recorded by NMR. Several different molecules can be caged: The photocage can be attached to RNA. Then, a modified photolabile nucleotide can be placed at strategic positions of a target RNA whose folding properties is to be studied. The photocage can also be attached to a ligand: if folding is dependent on ligand binding then the ligand can be modified to carry a photosensitive unit whose degradation allows binding to RNA. In this thesis, an alternative method for photocaging is introduced. Here, metal ions essential for folding of the RNA are photocaged using the photolabile chelating agent Dimethyl-nitrophen (DMN). Photolysis of DMNr releases the metal ion, thereby RNA folding is initiated. In the rapid-mixing technique, one of (several) components required for proper folding of the RNA is rapidly injected into an NMR sample in situ by the use of a pneumatic injection device. ...
Die Identifizierung neuartiger Verbindungsklassen für ein pharmakologisches Zielsystem ist eine fordernde Aufgabe für die frühe präklinische Forschung, insbesondere wenn bereits vorherige umfangreiche Studien durchgeführt und viele Leitstrukturserien gefunden wurden. In dieser Arbeit konnte gezeigt werden, dass Scaffold Hopping durch Methoden des Virtual Screenings auch für Systeme möglich ist, für die bereits eine Vielzahl von Referenzsubstanzen beschrieben ist und somit wenig freier chemischer Raum für Innovation zur Verfügung steht. Als Beispielsystem wurde die GlycinB-Bindungsstelle der NR1-Untereinheit des NMDA-Rezeptors betrachtet. Verschiedene zwei- und dreidimensionale Techniken des Virtual Screenings wurden einer umfangreichen retrospektiven Validierung unterworfen. Zur Durchführung der prospektiven Virtual-Screening-Studie wurde eine automatisierte in silico Plattform entwickelt, die 8,9 Millionen käufliche Substanzen aus 46 Substanzkatalogen von 33 verschiedenen Anbietern sammelte, um etwa 5 Millionen unterschiedliche Moleküle in zweidimensionaler Darstellung aufzuarbeiten. Diese Menge an Substanzen stellt den größten Teil der zurzeit kommerziell verfügbaren chemischen Verbindungen, also den „verfügbaren chemischen Raum“ dar. Anhand der retrospektiv validierten Virtual Screening Techniken konnten in einer prospektiven Suche 21 GlycinB-Antagonisten mit neuartigen, d.h. für GlycinB noch unbeschriebenen Scaffolds gefunden werden. Ausgehend von drei dieser Virtual Screening Hits wurden 53 weitere Verbindungen mit insgesamt fünf unterschiedlichen neuartigen Scaffolds und einem gemeinsamen Azo-Motiv identifiziert. Die Struktur-Wirkungsbeziehungen dieser fünf chemischen Serien wurden charakterisiert. Das Ergebnis dieser Arbeit zeigt eindeutig, dass es lohnend ist, alle vorhandenen Methoden auszuschöpfen, da sich die validierten Methoden komplementär zueinander verhielten und kein Virtual Screening Hit von mehr als einer Technik gefunden wurde. Die Flexibilität von Proteinen als Antwort auf die Bindung unterschiedlicher Liganden stellt ein bislang ungelöstes chemieinformatisches Problem dar, welches auch grundlegende pharmakologische Bedeutung hat. So verursachen z.B. bei NMDA/GlycinB agonistische Liganden eine Konformationsänderung des Rezeptors. Diese ruft dann eine direkte funktionale Antwort in Form der Öffnung des Ionenkanals hervor. Auch der Bindungsmodus der Antagonisten von GlycinB ist trotz Vorhandenseins von zwei Kristallstrukturen und mehreren Hundert zum Teil hochaffiner Referenzstrukturen zum großen Teil ungeklärt. Im zweiten Teil dieser Arbeit wurde ein auf Moleküldynamiksimulationen basierendes Verfahren entwickelt, welches flexible Aminosäurereste im Rezeptor und damit induzierbare Bewegungen des Proteinrückgrates bestimmt. Die so identifizierten Reste wurden dann in einem erweiterten Verfahren des Induced-Fit-Dockings als explizit flexibel betrachtet. Hierdurch war die Berechnung verschiedener Bindungsmodi von Antagonisten möglich, die aufgrund ihrer Form und Größe nicht in die verfügbaren Kristallstrukturen von GlycinB passten. Diese benötigten somit einen Induced-Fit-Effekt des Rezeptors, um eine Bindung einzugehen. Für die im ersten Teil dieser Arbeit identifizierten Azo-Liganden wurde auf Basis dieser Methode ein gemeinsamer Bindungsmodus vorgeschlagen. Ebenso konnte anhand der Methodik eine Aussage über die funktionale Auswirkung der Proteinflexibilität beim Übergang vom antagonistischen zum agonistischen Rezeptorzustand von GlycinB getroffen werden. Ein großes Problem aktueller Dockingverfahren ist die mangelnde Verfügbarkeit von Scoringfunktionen, welche die tatsächliche biologische Bindungsaffinität eines Liganden berechnen. Hier wurde ein Verfahren für das Zielsystem GlycinB gezeigt, welches aufgrund der Berechnung des thermodynamischen Entropie- und Enthalpiegewinns durch Verdrängung von hydrophob eingeschlossenen Wasser aus der Bindungsstelle durch den Liganden eine Aussage über dessen zu erwartende Bindungsaffinität trifft. Dieses neuartige Scoringsystem wurde auf die im Virtual Screening identifizierten Serie von Azo-Liganden angewandt und verfügte über eine im Vergleich zu klassischen Scoringfunktionen des Molecular Dockings verbesserte Vorhersagekraft der biologischen Bindungsaffinität.
"The whole is more than the sum of its parts." This idea has been brought forward by psychologists such as Max Wertheimer who formulated Gestalt laws that describe our perception. One law is that of collinearity: elements that correspond in their local orientation to their global axis of alignment form a collinear line, compared to a noncollinear line where local and global orientations are orthogonal. Psychophysical studies revealed a perceptual advantage for collinear over non-collinear stimulus context. It was suggested that this behavioral finding could be related to underlying neuronal mechanisms already in the primary visual cortex (V1). Studies have shown that neurons in V1 are linked according to a common fate: cells responding to collinearly aligned contours are predominantly interconnected by anisotropic long-range lateral connections. In the cat, the same holds true for visual interhemispheric connections. In the present study we aimed to test how the perceptual advantage of a collinear line is reflected in the anatomical properties within or between the two primary visual cortices. We applied two neurophysiological methods, electrode and optical recording, and reversibly deactivated the topographically corresponding contralateral region by cooling in eight anesthetized cats. In electrophysiology experiments our results revealed that influences by stimulus context significantly depend on a unit’s orientation preference. Vertical preferring units had on average a higher spike rate for collinear over non-collinear context. Horizontal preferring units showed the opposite result. Optical imaging experiments confirmed these findings for cortical areas assigned to vertical orientation preference. Further, when deactivating the contralateral region the spike rate for horizontal preferring units in the intact hemisphere significantly decreased in response to a collinear stimulus context. Most of the optical imaging experiments revealed a decrease in cortical activity in response to either stimulus context crossing the vertical midline. In conclusion, our results support the notion that modulating influences from stimulus context can be quite variable. We suggest that the kind of influence may depend on a cell’s orientation preference. The perceptual advantage of a collinear line as one of the Gestalt laws proposes is not uniformly represented in the activity of individual cells in V1. However, it is likely that the combined activity of many V1 neurons serves to activate neurons further up the processing stream which eventually leads to the perceptual phenomenon.
Im Rahmen dieser Arbeit wurden die 5´- und 3´-Enden 23 ausgewählter Transkripte des halophilen Archaeons Halobacterium salinarum bestimmt. Die Daten wurden dazu verwendet, die Längen der untranslatierten Bereiche zu ermitteln, Konsensussequenzen für die Transkriptionsinitiation und -termination abzuleiten und die Rolle haloarchaealer UTRs bei der Translationsinitiation und -regulation zu untersuchen. Der experimentelle Ansatz wurde mit einer bioinformatischen Analyse des Genoms von H. salinarum vervollständigt. Dabei konnten die Konsensussequenzen der basalen Promotorelemente genauer definiert werden und ein neues Promotorelement konnte entdeckt werden. Weiterhin wurde ein Konsensusmotiv gefunden, welches wahrscheinlich für die Termination der Transkription wichtig ist. Alle 23 analysierten Transkripte hatten eine 3´-UTR mit einer durchschnittlichen Länge von 48 Nukleotiden und ihre 3´-Enden waren nicht posttranskriptionell modifiziert. Die experimentellen Ergebnisse und bioinformatischen Daten ergaben, dass die Mehrheit der haloarchaealen Transkripte keine 5´-UTR besitzt. Die meisten Transkripte mit 5´-UTRs enthielten unerwarteterweise keine Shine-Dalgarno (SD)-Sequenz. Die Analyse des H. salinarum Genoms machte deutlich, dass weniger als 10% aller Gene eine SD Sequenz vorrausgeht und dass sogar bei Genen, die distal im Operon liegen, meistens keine SD-Sequenz zu finden ist. Weiterhin wurde der Einfluss einer ausgewählten 5´-UTR ohne SD-Sequenz und einer 3´-UTR auf die Transkriptstabilität und die Translationseffizienz untersucht. Das Transkript mit 5´ UTR ohne SD-Sequenz wurde in H. salinarum effizient translatiert. In einer Studie an H. volcanii konnte das Gleiche für verschiedene 5´ UTRs ohne SD-Sequenz in Verbindung mit einem Reportergen gezeigt werden (Brenneis et al., 2007). An diesen Transkripten kann die Translation also nicht über den „bakteriellen“ Mechansimus durch Basenpaarung mit dem 3´-Ende der 16S rRNA initiiert werden. Im weiteren Verlauf der Arbeit wurde untersucht, ob es sich bei dem Mechanismus der Translationsinitiation an Transkripten mit 5´-UTR ohne SD-Sequenz um einen Scanning-Mechanismus wie bei Eukaryonten oder um einen neuen Mechanismus handelt. Für die Experimente wurde das zuvor erwähnte Reportergensystem aus H. volcanii verwendet. Neben AUG wurden GUG und UUG effizient als Startkodons an einem Transkript mit 5´-UTR genutzt, wohingegen AUG das einzige funktionierende Startkodon bei einem Transkript ohne 5´-UTR war. Verschiedene Deletionsversionen der 20 Nukleotide langen 5´-UTR wurden im Gegensatz zur kompletten 5´-UTR nur sehr ineffizient translatiert. Das Einfügen zusätzlicher AUGs stromaufwärts des natürlichen Startkodons hatte keinen Einfluss auf die Translationseffizienz am internen AUG. Ein zusätzliches AUG am 5´-Ende im gleichen Leseraster des natürlichen AUGs führte zur gleichzeitigen Nutzung beider Startkodons auf derselben mRNA. Eine stabile Haarnadelstruktur am 5´-Ende inhibierte die Translation nur am ersten AUG und hatte keinen Einfluss auf die Translationseffizienz am internen AUG. Zusammenfassend konnte durch die erhaltenen Ergebnisse ausgeschlossen werden, dass der Mechanismus der Translationsinitiation an Transkripten mit 5´-UTR ohne SD-Sequenz ein Scanning-Mechansimus wie bei Eukaryonten ist. Neben der Translationsinitiation an Transkripten ohne 5´-UTR und an Transkripten mit SD-Sequenz existiert also noch ein dritter, bis jetzt unbekannter Mechansimus zur Translationsinitiation in Haloarchaea. In initialen Versuchen zur genaueren Charakterisierung der drei verschiedenen Translationsinitiationsmechanismen, wurden Konstrukte hergestellt, bei denen alle drei verschiedenen Startkodons auf einer mRNA vorhanden sind. Es wurden erste Hinweise darauf gefunden, dass Stressbedingungen einen Einfluss auf die unterschiedlichen Initiationsmechansimen haben. Anders als erwartet führte die Mutation einer natürlichen SD-Sequenz nicht zu einer verminderten Translationseffizienz am entsprechenden Startkodon. Die Bestimmung des 5´-Endes eines Transkripts mit in silico vorhergesagter SD-Sequenz offenbarte, dass das Transkript keine 5´-UTR und somit auch keine SD-Sequenz besaß. Beide Ergebnisse machen deutlich, dass SD-Sequenzen eine noch viel geringere Rolle bei der haloarchaealen Translationsinitiation spielen, als anhand der Sequenz und UTR Analysen angenommen werden konnte.
Photosystem (PS) I is a huge membrane protein complex which coordinates around 200 co-factors. Upon light excitation a charge separation at the PS I reaction centre is induced which leads to an electron transport across the thylakoid membrane and the generation of redox equivalents needed for several biochemical reactions, e.g. the synthesis of sugars. For higher plants and cyanobacteria the crystal structure of PS I complexes were resolved to resolutions of 4.4 Å and 2.5 Å. Furthermore, supramolecular structures of PS I of eukaryotic algae, mainly of the green line, were obtained recently. However, up to now, no structure of diatoms is available yet. Diatoms are key players in global primary production and derived from a secondary endosymbiosis event. Their chloroplasts are surrounded by four envelope membranes and their thylakoids are evenly arranged in bands of three, i.e. no separation in grana and stroma regions is apparent. In this thesis a protocol was developed to isolate a functional PS I complex of diatoms which can be used for structural analysis by transmissional electron microscopy (TEM). A photosystem I-fucoxanthin chlorophyll protein (PS I-FCP) complex was isolated from the pennate diatom Phaeodactylum tricornutum by ion exchange chromatography. Spectroscopic analysis proved that bound Fcp polypeptides function as a light-harvesting complex. An active light energy transfer from Fcp associated pigments, Chl c and fucoxanthin, towards the PS I core was proven by fluorescence spectroscopy. Oxidised minus reduced difference spectroscopy evidenced the activity of the PS I reaction centre P700 and yielded a chlorophyll a/P700 ratio of approximately 200:1. These data indicate that the isolated PS I-FCP complex exceeds the PS I cores from cyanobacteria and higher plants in the numbers of chlorophyll a molecules. Because of the strict conservation of PS I cores among organisms the additional 100 chlorophyll a molecules must either be coordinated by Fcps or function as linker molecules between the Fcp antenna and the PS I core as shown for the PS I-LHC I complex of higher plants. To tell something about the structural organisation, the PS I-FCP complex was compared with its cyanobacterial and higher plant counterparts. Whereas cyanobacterial PS I cores aggregate to trimers, usually without associated antennae, higher plant PS I is a monomer and binds additionally two LHC I heterodimers. BN-PAGE and gel filtration experiments showed that also diatoms contain PS I monomers associated with Fcps as light-harvesting antenna. First TEM studies evidenced these observations. Negatively stained PS I-FCP particles had an increased size compared to PS I cores of other organisms. No PS I trimers or higher oligomers have been found. The calculated diameter and shape of the particles correspond to PS I-LHC I particles obtained from green algae, which also comprise of a higher number of LHC I polypeptides compared to the higher plant x-ray structure. Additionally, the analysis of polypeptides indicates that the PS I associated Fcps differ from the free Fcp pool and also from Fcps of a PS II enriched fraction. The assumption that diatoms harbour just one Fcp antenna that serve both Photosystems equally seems to be wrong. To further study the association of Fcps with the two Photosystems, both complexes plus the free FCP complexes were isolated from the centric diatom Cyclotella meneghiniana. Because of the availability of antibodies directed against specific Fcp polypeptides of Cyclotella the PS I-FCP complex of Phaeodactylum could not be used. A trimeric FCP complex, FCPa, and a higher FCP oligomer, FCPb, have already been described for C. meneghiniana. The latter is assumed to be composed of only Fcp5, whereas the FCPa contains Fcp2 and Fcp6. Biochemical and spectroscopical evidences revealed a different subset of associated Fcp polypeptides within the isolated photosystem complexes. Whereas the PS II associated Fcp antenna resembles FCPa, at least three different Fcp polypeptides are associated with PS I. By re-solubilisation of the PS I complex and a further purification step Fcp polypeptides were partially removed from PS I and both fractions were analysed again by biochemical and spectroscopical means, as well as by HPLC. Thereby Fcp4 and a so far undescribed 17 kDa Fcp were found to be strongly coupled to PS I, whereas another Fcp, presumably Fcp5, is only loosely bound to the PS I core. Thus an association of FCPb and PS I is assumed.
Background: In this interdisciplinary project, the biological effects of heavy ions are compared to those of X-rays using tissue slice culture preparations from rodents and humans. Advantages of this biological model are the conservation of an organotypic environment and the independency from genetic immortalization strategies used to generate cell lines. Its open access allows easy treatment and observation via live-imaging microscopy. Materials and methods: Rat brains and human brain tumor tissue are cut into 300 micro m thick tissue slices. These slices are cultivated using a membrane-based culture system and kept in an incubator at 37°C until treatment. The slices are treated with X-rays at the radiation facility of the University Hospital in Frankfurt at doses of up to 40 Gy. The heavy ion irradiations were performed at the UNILAC facility at GSI with different ions of 11.4 A MeV and fluences ranging from 0.5–10 x 106 particles/cm². Using 3D-confocal microscopy, cell-death and immune cell activation of the irradiated slices are analyzed. Planning of the irradiation experiments is done with simulation programs developed at GSI and FIAS. Results: After receiving a single application of either X-rays or heavy ions, slices were kept in culture for up to 9d post irradiation. DNA damage was visualized using gamma H2AXstaining. Here, a dose-dependent increase and time-dependent decrease could clearly be observed for the X-ray irradiation. Slices irradiated with heavy ions showed less gamma H2AX-positive cells distributed evenly throughout the slice, even though particles were calculated to penetrate only 90–100 micro m into the slice. Conclusions: Single irradiations of brain tissue, even at high doses of 40 Gy, will result neither in tissue damage visible on a macroscopic level nor necrosis. This is in line with the view that the brain is highly radio-resistant. However, DNA damage can be detected very well in tissue slices using gamma H2AX-immuno staining. Thus, slice cultures are an excellent tool to study radiation-induced damage and repair mechanisms in living tissues.
This article discusses the divergent status of the two particles lé and lá in the grammar of Konkomba, a Gur language (Niger-Congo) of the Gurma subgroup. While previous studies claim that both particles are focus markers, this author argues that only the particle lá should be analyzed as a pure pragmatic device. Distributional studies suggest that the use of particle lé, on the other hand, is only required under specific focus conditions, and primarily represents a syntactic device.
Die drei Bereiche, die hier verglichen werden sollen, entsprechen in etwa der überkommenen Trias von Literatur, Musik und bildender Kunst, einer Gliederung, die im Medienzeitalters mit Videos, CDs, Installationen oder Happenings eigentlich obsolet ist. Allerdings geht es hier nur um die Eigenart der Zeichensysteme, auf denen die verschiedenen Bereiche beruhen, nicht um die Werke, die dadurch möglich werden, obgleich natürlich auch die Kunstwerke im emphatischen Sinn, die bedeutenden und die banalen, die großen und die misslungenen Gestaltungen nur möglich und verstehbar sind aufgrund der Zeichen, auf denen sie beruhen.
The main tenet of the present paper is the thesis that nominalization – like other cases of derivational morphology – is an essentially lexical phenomenon with well defined syntactic (and semantic) conditions and consequences. More specifically, it will be argued that the relation between a verb and the noun derived from it is subject to both systematic and idiosyncratic conditions with respect to lexical as well as syntactic aspects.
Local interactions between particles of a collection causes all particles to reorganize in new positions. The purpose of this paper is to construct an energy-based model of self-organizing subgroups, which describes the behavior of singular local moves of a particle. The present paper extends the Hegselmann-Krause model on consensus dynamics, where agents simultaneously move to the barycenter of all agents in an epsilon neighborhood. Mit dem Ende der unmittelbaren Zeitzeugengeneration rückt besonders für die regionalen und lokalen Forschungen zur Shoa und zur Herrschaft des Nationalsozialismus insgesamt eine bisher eher vernachlässigte Quellengattung in den Blickpunkt: die Berichte und Erinnerungen der so genannten „zweiten Generation der Überlebenden“. Die bisher erschienene Literatur konzentrierte sich weitgehend auf die therapeutische Bearbeitung von deren Traumata. Die Berichte derer, die als kleine Kinder überlebten, und der Nachgeborenen als historische Quellen blieben dagegen nahezu unerschlossen. Es ist dies selbstverständlich auch den besonderen Schwierigkeiten dieser Erinnerungen und Aussagen geschuldet, die oft eine Widerspiegelung des Schweigens sind. Weniger das Ausgesprochene, sondern das Ungesagte und Unsagbare prägt das Selbstverständnis dieser Generation. Fragmente dieser Geschichte wirken brüchig bis in die Gegenwart.
