Die Weimarer Beiträge - seit ihrer Einstellung durch den Aufbauverlag 1991 vom Passagen Verlag herausgegeben - ist eine der renommiertesten Literatur- und Kulturzeitschriften der ehemaligen DDR. Durch ihren interdisziplinären Ansatz, der auch allgemeine kulturelle, ästhetische und politische Überlegungen einbezieht, trägt sie zu einer Einbindung der deutschsprachigen Kulturwissenschaften in die internationale Diskussion bei.
The aim of this introductory article [to the volume of the same title], firstly, is to recapitulate the basic principles of Bakhtin’s initial theory as formulated in “Forms of Time and of the Chronotope in the Novel: Notes toward a Historical Poetics” (henceforth FTC) and “The Bildungsroman and its Significance in the History of Realism (Toward a Historic Typology of the Novel)” (henceforth BSHR). Subsequently, we present some relevant elaborations of Bakhtin’s initial concept and a number of applications of chronotopic analysis, closing our state of the art by outlining two perspectives for further investigation. Some of the issues which we touch upon receive more detailed treatment in other contributions to this volume. Others may offer perspectives for future Bakhtin scholarship.
Das Exil war sein Schicksal. Davon ist er zeit seines Lebens nicht losgekommen. Hans Sahl, aus Dresden gebürtig, gehörte zu jenen brillanten Intellektuellen, die bereits im geistigen Leben der Weimarer Republik eine Rolle spielten, vor den Nazis fliehen mußten und als ungeliebte Heimkehrer in der Bundesrepublik lange vergessen blieben. Erst in seinen letzten Lebensjahren, die der fast erblindete Autor in Tübingen verbrachte, wurde er mit Preisen überschüttet, wurden seine Gedichte und Romane neuaufgelegt. “Wir sind die Letzten“, hieß eine Gedichtsammlung, die 1941 erschien, in der sich dieser „Trödler des Unbegreiflichen“, wie er sich nannte, als Moralist und Wahrheitsfanatiker von hohen Graden zu erkennen gab.
This edited volume is the first scholarly tome exclusively dedicated to Mikhail Bakhtin’s theory of the literary chronotope. This concept, initially developed in the 1930s and used as a frame of reference throughout Bakhtin’s own writings, has been highly influential in literary studies. After an extensive introduction that serves as a ‘state of the art’, the volume is divided into four main parts: Philosophical Reflections, Relevance of the Chronotope for Literary History, Chronotopical Readings and Some Perspectives for Literary Theory. These thematic categories contain contributions by well-established Bakhtin specialists such as Gary Saul Morson and Michael Holquist, as well as a number of essays by scholars who have published on this subject before. Together the papers in this volume explore the implications of Bakhtin’s concept of the chronotope for a variety of theoretical topics such as literary imagination, polysystem theory and literary adaptation; for modern views on literary history ranging from the hellenistic romance to nineteenth-century realism; and for analyses of well-known novelists and poets as diverse as Milton, Fielding, Dickinson, Dostoevsky, Papadiamandis and DeLillo
Although autism spectrum disorders (ASDs) have a substantial genetic basis, most of the known genetic risk has been traced to rare variants, principally copy number variants (CNVs). To identify common risk variation, the Autism Genome Project (AGP) Consortium genotyped 1558 rigorously defined ASD families for 1 million single-nucleotide polymorphisms (SNPs) and analyzed these SNP genotypes for association with ASD. In one of four primary association analyses, the association signal for marker rs4141463, located within MACROD2, crossed the genome-wide association significance threshold of P < 5 × 10−8. When a smaller replication sample was analyzed, the risk allele at rs4141463 was again over-transmitted; yet, consistent with the winner's curse, its effect size in the replication sample was much smaller; and, for the combined samples, the association signal barely fell below the P < 5 × 10−8 threshold. Exploratory analyses of phenotypic subtypes yielded no significant associations after correction for multiple testing. They did, however, yield strong signals within several genes, KIAA0564, PLD5, POU6F2, ST8SIA2 and TAF1C.

Das Paar Lou/ Nietzsche hat die Phantasie späterer Generationen beflügelt, dieses Zuendedenken und –leben nicht erfüllter Wünsche und Sehnsüchte hat das Kolorit beider Erinnerung wesentlich überzeichnet.
Der IT-Industrieverband Bitkom findet die Debatte um Netzneutralität nicht so gelungen und möchte lieber über etwas anderes reden: Deutsche IT-Branche ringt um Position zur Netzneutralität. Dass die Deutsche Telekom bei Bitkom eines der einflußreichsten Mitglieder ist, wenn nicht sogar das einflußreichste, dürfte damit sicher überhaupt nichts zu tun haben.
Background: The apoptosis-inducing serine protease granzyme B (GrB) is an important factor contributing to lysis of target cells by cytotoxic lymphocytes. Expression of enzymatically active GrB in recombinant form is a prerequisite for functional analysis and application of GrB for therapeutic purposes. Methods and Findings: We investigated the influence of bacterial maltose-binding protein (MBP) fused to GrB via a synthetic furin recognition motif on the expression of the MBP fusion protein also containing an N-terminal alpha-factor signal peptide in the yeast Pichia pastoris. MBP markedly enhanced the amount of GrB secreted into culture supernatant, which was not the case when GrB was fused to GST. MBP-GrB fusion protein was cleaved during secretion by an endogenous furin-like proteolytic activity in vivo, liberating enzymatically active GrB without the need of subsequent in vitro processing. Similar results were obtained upon expression of a recombinant fragment of the ErbB2/HER2 receptor protein or GST as MBP fusions. Conclusions: Our results demonstrate that combination of MBP as a solubility enhancer with specific in vivo cleavage augments secretion of processed and functionally active proteins from yeast. This strategy may be generally applicable to improve folding and increase yields of recombinant proteins.
Background: Early inner ear development requires the strict regulation of cell proliferation, survival, migration and differentiation, coordinated by the concerted action of extrinsic and intrinsic factors. Deregulation of these processes is associated with embryonic malformations and deafness. We have shown that insulin-like growth factor I (IGF-I) plays a key role in embryonic and postnatal otic development by triggering the activation of intracellular lipid and protein kinases. RAF kinases are serine/threonine kinases that regulate the highly conserved RAS-RAF-MEK-ERK signaling cascade involved in transducing the signals from extracellular growth factors to the nucleus. However, the regulation of RAF kinase activity by growth factors during development is complex and still not fully understood.
Methodology/Principal Findings: By using a combination of qRT-PCR, Western blotting, immunohistochemistry and in situ hybridization, we show that C-RAF and B-RAF are expressed during the early development of the chicken inner ear in specific spatiotemporal patterns. Moreover, later in development B-RAF expression is associated to hair cells in the sensory patches. Experiments in ex vivo cultures of otic vesicle explants demonstrate that the influence of IGF-I on proliferation but not survival depends on RAF kinase activating the MEK-ERK phosphorylation cascade. With the specific RAF inhibitor Sorafenib, we show that blocking RAF activity in organotypic cultures increases apoptosis and diminishes the rate of cell proliferation in the otic epithelia, as well as severely impairing neurogenesis of the acoustic-vestibular ganglion (AVG) and neuron maturation.
Conclusions/Significance: We conclude that RAF kinase activity is essential to establish the balance between cell proliferation and death in neuroepithelial otic precursors, and for otic neuron differentiation and axonal growth at the AVG.
Background: Falciparum Malaria, an infectious disease caused by the apicomplexan parasite Plasmodium falciparum, is among the leading causes of death and morbidity attributable to infectious diseases worldwide. In Gabon, Central Africa, one out of four inpatients have severe malarial anemia (SMA), a life-threatening complication if left untreated. Emerging drug resistant parasites might aggravate the situation. This case control study investigates biomarkers of enhanced hemolysis in hospitalized children with either SMA or mild malaria (MM). Methods and Findings: Ninety-one children were included, thereof 39 SMA patients. Strict inclusion criteria were chosen to exclude other causes of anemia. At diagnosis, erythrophagocytosis (a direct marker for extravascular hemolysis, EVH) was enhanced in SMA compared to MM patients (5.0 arbitrary units (AU) (interquartile range (IR): 2.2–9.6) vs. 2.1 AU (IR: 1.3–3.9), p<0.01). Furthermore, indirect markers for EVH, (i.e. serum neopterin levels, spleen size enlargement and monocyte pigment) were significantly increased in SMA patients. Markers for erythrocyte ageing, such as CD35 (complement receptor 1), CD55 (decay acceleration factor) and phosphatidylserine exposure (annexin-V-binding) were investigated by flow cytometry. In SMA patients, levels of CD35 and CD55 on the red blood cell surface were decreased and erythrocyte removal markers were increased when compared to MM or reconvalescent patients. Additionally, intravascular hemolysis (IVH) was quantified using several indirect markers (LDH, alpha-HBDH, haptoglobin and hemopexin), which all showed elevated IVH in SMA. The presence of both IVH and EVH predicted the need for blood transfusion during antimalarial treatment (odds ratio 61.5, 95% confidence interval (CI): 8.9–427). Interestingly, this subpopulation is characterized by a significantly lowered reticulocyte production index (RPI, p<0.05). Conclusions: Our results show the multifactorial pathophysiology of SMA, whereby EVH and IVH play a particularly important role. We propose a model where removal of infected and non-infected erythrocytes of all ages (including reticulocytes) by EVH and IVH is a main mechanism of SMA. Further studies are underway to investigate the mechanism and extent of reticulocyte removal to identify possible interventions to reduce the risk of SMA development.
Recombinase-mediated cassette exchange (RMCE) exploits the possibility to unidirectionally exchange any genetic material flanked by heterotypic recombinase recognition sites (RRS) with target sites in the genome. Due to a limited number of available pre-fabricated target sites, RMCE in mouse embryonic stem (ES) cells has not been tapped to its full potential to date. Here, we introduce a universal system, which allows the targeted insertion of any given transcriptional unit into 85 742 previously annotated retroviral conditional gene trap insertions, representing 7013 independent genes in mouse ES cells, by RMCE. This system can be used to express any given cDNA under the control of endogenous trapped promoters in vivo, as well as for the generation of transposon ‘launch pads’ for chromosomal region-specific ‘Sleeping Beauty’ insertional mutagenesis. Moreover, transcription of the gene-of-interest is only activated upon Cre-recombinase activity, a feature that adds conditionality to this expression system, which is demonstrated in vivo. The use of the RMCE system presented in this work requires one single-cloning step followed by one overnight gateway clonase reaction and subsequent cassette exchange in ES cells with efficiencies of 40% in average.
We solved the crystal structure of a novel type of c-ring isolated from Bacillus pseudofirmus OF4 at 2.5 Å, revealing a cylinder with a tridecameric stoichiometry, a central pore, and an overall shape that is distinct from those reported thus far. Within the groove of two neighboring c-subunits, the conserved glutamate of the outer helix shares the proton with a bound water molecule which itself is coordinated by three other amino acids of outer helices. Although none of the inner helices contributes to ion binding and the glutamate has no other hydrogen bonding partner than the water oxygen, the site remains in a stable, ion-locked conformation that represents the functional state present at the c-ring/membrane interface during rotation. This structure reveals a new, third type of ion coordination in ATP synthases. It appears in the ion binding site of an alkaliphile in which it represents a finely tuned adaptation of the proton affinity during the reaction cycle. Mediengestützter Deutschunterricht im türkischen universitären Bereich : eine Bestandsaufnahme

A trend in nature of a permanent increase towards multimedia lifestyle has arisen in all stratas of the society. Thus, rather than using written course-books, publishing houses prefer to encourage use of multimedia which are dependent to course-book or which are independent of course-book and language learners prefer to learn with multimedia. Thus it is encouraged that courses are supported in that manner. This study aims to examine scope and limits of computer aided German teaching which is flourishing as a foreign language within Turkey university education recently. This study has been applied in preparatory classes of departments which provide four-year education. Results of a survey on use of multimedia dependent on course-book or independent of course-book within courses within Turkey university education has been given within scope of this study. Evidences on competence of German teachers and learners in use of multimedia has been given and have been visualized through use of graphics. Problems of multimedia aided German courses and solutions offers will be submitted.
Nach den Worten des Bundesverfassungsgerichts ist die Gewaltenteilung ein tragendes Organisationsprinzip des Grundgesetzes (BVerfGE 3, S. 225 ff. [247]). Keine Gewalt darf ein von der Verfassung nicht vorgesehenes Übergewicht über die andere Gewalt erhalten (BVerfGE 9, S. 268 ff. [279]). Wo in der deutschen Verfassungswirklichkeit im Verhältnis zwischen Exekutive und Judikative solche Organisationsstrukturen von zureichender Wirkungskraft zu finden sind, stellt das Bundesverfassungsgericht nicht klar. Die allein mit rechtswissenschaftlichen Instrumentarien nicht zu klärende Frage, ob die Bundesrepublik Deutschland bereits Organisationsstrukturen besitzt, die ein psychosoziales Übergewicht der Exekutive über die Rechtsprechung ausschließen, begegnet bei näherer Betrachtung der Verfassungswirklichkeit in Deutschland (Kapitel III., Schwerpunkt der Dissertation) erheblichen Zweifeln. Die nach dem Ende des 2. Weltkriegs vorgefundene und bis heute in ihrer Grundstruktur unveränderte Verwaltung der Dritten Gewalt durch die Exekutive entspricht nicht den Zielen des Gewaltenteilungsprinzips. Ein Prinzip ist eine Einsicht, ein Ziel und eine Handlungsregel, die methodisch am Anfang eines theoretischen Aufbaus oder Systems von Handlungsorientierungen steht. Montesquieu umschreibt die seinem Gewaltenteilungsprinzip zu Grunde liegende anthropologische Einsicht mit den Worten: „Eine ewige Erfahrung lehrt jedoch, dass jeder Mensch, der Macht hat, dazu getrieben wird, sie zu missbrauchen. Er geht immer weiter, bis er an Grenzen stößt. Wer hätte das gedacht: Sogar die Tugend hat Grenzen nötig“. Die deutsche Verfassungswirklichkeit der Rechtsprechung kennt keine Schutzgrenzen gegenüber der Exekutive. Es gibt keine Organisationsstrukturen, die subtile Übergriffe der vollziehenden Gewalt auf die Rechtsprechung von vornherein objektiv unmöglich machten. Deutschland begnügt sich mit Appellen an die mit der Justizverwaltung betrauten Regierungsmitglieder (i.d.R. Justizminister) und an die ihnen nachgeordneten Staatsorgane (z.B. Gerichtspräsidenten); sie hofft auf deren Rechtstreue und Augenmaß. Deutschland folgt dem Hoffnungsprinzip, nicht dem Gewaltenteilungsprinzip. Das im Grundgesetz festgeschriebene Prinzip der Gewaltenteilung organisatorisch umzusetzen, ist ein Ziel und eine Handlungsregel und damit eine vom Grundgesetz gestellte Aufgabe (a.M. BVerfGE 55, S. 372 ff. [388]). Die der Dreiteilung der Staatsgewalten (Art. 20 II 2 GG) vorausgehende anthropologische Einsicht Montesquieus ist überdies einer der bei der Auslegung des Grundgesetzes anzulegenden Maßstäbe. Der Wortlaut des Art. 92 erster Halbsatz GG ist die an die Spitze des Abschnitts „IX. Die Rechtsprechung“ gestellte Bestätigung der Grundentscheidung des Art. 20 II 2 GG für die Gewaltenteilung. Er bringt den Willen des Grundgesetzes zum Ausdruck, dass in der Verfassungswirklichkeit Deutschlands die rechtsprechende Gewalt in einer organisatorisch gleich wirkungsvollen Weise den Richtern anvertraut sein möge wie die gesetzgebende Gewalt den Parlamentsabgeordneten.
Das Neuroblastom ist ein Tumor, der sich von sympathoadrenergen Vorläuferzellen ableitet und die häufigste solide Krebsform im Kindesalter darstellt. Das breite klinische Spektrum dieses Tumors, das von spontaner Regression zu fataler Progression reicht, spiegelt die außerordentliche biologische und genetische Heterogenität dieses Tumors wider. Polyploidie und Genexpressionsanalysen werden auf klinischer Seite zur Risiko- und Therapieeinschätzung eingesetzt. Genomweite Screeninganalysen identifizierten den Transkriptionsfaktor Phox2b als erstes Prädispositionsgen für NB. Frühere Arbeiten haben gezeigt, dass Phox2b absolut essentiell für die Entstehung aller Ganglien des autonomen NS aus Neuralleistenzellen ist. Ziel der Untersuchungen dieser Arbeit war es, die Auswirkungen der NB-assoziierten Phox2b-Mutationen auf Proliferations- und Differenzierungsverhalten sympathoadrenerger Vorläuferzellen zu untersuchen. Hierzu wurden paravertebrale, sympathische Grenzstränge aus Huhnembryonen des Embryonaltags 7 präpariert, dissoziiert und mit Expressionsplasmiden für Phox2bwt und NB-Phox2b-Mutationen transfiziert. Nach zwei Tagen in Kultur wurden mit molekularbiologische Methoden Veränderungen des Proliferations-und Differenzierungsverhalten untersucht. Die Analyse des Proliferationsverhaltens transfizierter Neurone mit BrdU-Proliferationsassays und Phospho-Histon-3-Antikörpern offenbarte einen stark antiproliferativen Effekt des Phox2bwt-Proteins, den die untersuchten NB-Phox2b-Mutationen nicht aufwiesen. NB-Phox2b-Patienten sind heterozygot, d.h. Träger eines gesunden und eines mutierten Phox2b-Allels. Um die genetische Situation im NB-Patienten nachzuahmen, wurde mit spezifischen siRNAs das endogene Phox2b-Protein-Niveau herunter reguliert. NB-Phox2b-Mutationen mit mis- oder nonsense Mutation in der Homöodomäne zeigten unter Phox2b-Knockdown-Bedingungen einen proliferationsstimulierenden Effekt. Diese Experimente warfen die Frage auf, über welche Mechanismen Phox2b und NB-Phox2b-Mutationen Einfluss auf die Zellzykluskontrolle nehmen. Die quantitative Analyse der Expression bekannter Zellzyklusregulatoren wie Zyklin D1, D2 und D3 und der Zyklin-abhängigen Kinase-Inhibitoren p18, p21 und p57kip2 verlief ergebnislos. Die Überexpression des Zyklin-abhängigen Kinase-Inhibitors p27kip1 wirkte antiproliferativ auf Kulturen sympathoadrenerger Vorläuferzellen und das p27kip1-Epxressionsniveau korrelierte mit dem Phox2b-Proteinniveau. P27kip1 scheidet jedoch als alleiniger Vermittler des antiproliferativen Effekts von Phox2b aus, da die ebenfalls antiproliferativ wirkende Phox2-Homöodomäne keinen Einfluss auf das p27kip1-Expressionsniveau besitzt. Vielmehr wurde der bHLH-Transkriptionsfaktor Hand2 als Mediator der Proliferationseffekte von Phox2b identifiziert. Die Proliferation sympathoadrenerger Vorläuferzellen ist abhängig von der Hand2-Proteinmenge, und Hand2-Überexpression ist ausreichend, den antiproliferativen Effekt von Phox2b aufzuheben. Damit im Einklang geht der proliferationsstimulierende Effekt der Phox2bHDmut bei siRNA-vermitteltem Hand2-Knockdown verloren. Weiterhin führt Phox2b-Überexpression zu verringertem Hand2-Expressionsniveau und Phox2b-Knockdown zu vermehrter Hand2-Transkription. In Protein-Protein-Interaktionsexperimenten konnte eine direkte Bindung von Hand2 an Phox2bwt und die untersuchten NB-Phox2b-Mutanten nachgewiesen werden. Diese Ergebnisse zeigen, dass Hand2 in der Vermittlung der Phox2b-Proliferationseffekte auf transkriptioneller und posttranskriptioneller Ebene involviert ist. Der Wirkungsmechanismus dieser Phox2b-Varianten / Hand2-Komplexe konnte nicht endgültig geklärt werden und wird in Form verschiedener Modelle diskutiert. In NB-Patienten korreliert ein hohes Expressionsniveau von Differenzierungsmarkern mit mildem Krankheitsverlauf. Die Rolle von Phox2b als Schlüsselgen in der Entstehung und Differenzierung autonomer Neurone und die genetische Heterogenität des NB legen eine Funktion von NB-Phox2b-Mutationen auf den Differenzierungsstatus sympathoadrenerger Vorläuferzellen nahe. In quantitativen Analysen wurde die Expression von Phox2b-Zielgenen und in NB-Diagnostik involvierten Genen untersucht. Phox2bK155X, eine C-terminal trunkierte NB-Phox2b-Mutation, wirkte dominant-negativ auf die Expression der noradrenergen Markergene Th und Dbh, Tlx3 und die Neurotrophinrezeptoren trkA und p75, deren reduzierte Expression mit schlechter Prognose und damit aggressiven NB-Formen korreliert ist. Zusammenfassend konnte in dieser Arbeit gezeigt werden, das NB-Phox2b-Mutationen in sympathoadrenergen Vorläuferzellen, den potentiellen Tumorentstehungszellen des NB, proliferationsstimulierend wirken und zumindest die C-terminal trunkierte Phox2bK155X-Mutation zur Dedifferenzierung dieser Zellen führt. Hereditäre Mutationen in Phox2b könnten im Patienten nicht nur die terminale Differenzierung sympathischer Neurone stören, sondern auch durch eine verlängerte Phase der Neurogenese die Empfänglichkeit für weitere Tumor-initiierende Mutation erhöhen.
In dieser Arbeit wurde ein Verfahren entwickelt, mit dem die Entfaltung von Proteinen simuliert werden kann. Anhand der Simulation kann die Schmelztemperatur bestimmt und damit die Thermostabilität einer Struktur untersucht werden. Außerdem ist die Untersuchung struktureller Veränderungen möglich, die während der Entfaltung auftreten und letztendlich zum globalen Zerfall der Struktur führen. Die Bereiche, von denen der globale Zerfall der Struktur ausgeht, können bestimmt werden. Es wurde untersucht, inwieweit diese Entfaltungsregionen Strukturbereiche darstellen, deren Mutation die thermische Stabilität der Struktur beeinflusst. Das entwickelte Verfahren basiert auf der Anwendung von Methoden aus der Rigiditätstheorie. Die thermische Entfaltung der Strukturen wird über das sukzessive Aufbrechen nichtkovalenter Wechselwirkungen in den Netzwerken simuliert. An-sätze aus der Perkolations- und Netzwerktheorie werden verwendet, um die Rigidität und Flexibilität in den Netzwerken während der Entfaltung zu untersuchen. Diese unter dem Begriff der Analyse statischer Netzwerke (constraint network analysis, CNA) zusammengefassten Methoden wurden im ersten Teil dieser Arbeit auf einen Datensatz homologer meso- und thermophiler Proteine angewendet. Dabei wurde untersucht, ob die thermophile Anpassung tatsächlich über eine Rigidisierung der Struktur erfolgt. Außerdem wurde die Theorie der korrespondierenden Zustände getestet, die besagt, dass bei der thermophilen Anpassung trotz globaler Rigidisierung für die Bioaktivität wichtige flexible Bereiche konserviert sind. Mit Hilfe der CNA wurden Entfaltungsregionen der Proteine aus dem Datensatz bestimmt und mit Strukturbereichen verglichen, in die thermostabilisierende Mutationen eingeführt wurden. Außerdem wurde untersucht, inwieweit vorhergesagt werden kann, ob eine möglicherweise thermostabilisierende Mutation die Aktivität negativ beeinflusst. Für zwei Drittel der thermophilen Proteine aus dem Datensatz konnte eine höhere Thermostabilität vorhergesagt werden als für das entsprechende mesophile Protein. Es konnte zudem gezeigt werden, dass die thermophile Anpassung dieser Proteine tatsächlich über eine Rigidisierung der Struktur erfolgt. Offensichtlich werden bei der Anwendung der CNA implizit alle möglichen Mechanismen der thermophilen Anpassung berücksichtigt. Die für zwei Paare meso- und thermophiler Proteine vorhergesagten Entfaltungsregionen stimmten sehr gut mit Bereichen überein, in die thermostabilisierende Mutationen eingeführt wurden. Damit wurde gezeigt, dass die CNA hilfreich zur Unterstützung des Protein Engineering ist, da die thermische Stabilität einer Struktur abgeschätzt werden kann, andererseits Hinweise darauf gegeben werden, in welchen Bereichen der Struktur thermostabilisierende Mutationen eingeführt werden können. Anhand des Vergleichs mikroskopischer Stabilitäten homologer Proteine konnte gezeigt werden, dass die CNA ein Abschätzen des Effekts einer thermostabilisierenden Mutation auf die Aktivität erlaubt, was wiederum für den Einsatz der CNA zur Unterstützung des Protein Engineering spricht. Im zweiten Teil dieser Arbeit wurde die CNA auf eine Serie von Phytasen unterschiedlicher Thermostabilitäten angewendet. Da es sich bei den Phytase-Strukturen um Homologie-modelle handelte, die nicht direkt mit der Analyse statischer Netzwerke untersucht werden konnten, wurde eine ensemblebasierte CNA etabliert. Dazu wurden kurze MD-Simulationen zur Verbesserung der homologiemodellierten Strukturen durchgeführt, aus denen dann ein konformationelles Strukturensemble extrahiert wurde. Aus den konformationellen Ensembles werden Thermostabilitäten vorhergesagt. Bei der Vorhersage der Thermostabilitäten ergab sich eine bemerkenswert gute Übereinstimmung mit experimentell bestimmten relativen Halbwertszeittemperaturen. Bereiche mit hoher Entfaltungsregionwahrscheinlichkeit stimmen gut mit Regionen überein, in denen thermostabilisierende Mutationen experimentell eingeführt wurden. Diese Ergebnisse offenbaren, dass mit der Entwicklung der ensemblebasierten CNA die methodischen Grundlagen für den Einsatz des Verfahrens zur Unterstützung des Protein Engineering von Phytasen geschaffen wurden.
The thermodynamics of base pairing is of fundamental importance. Fluorinated base analogs are valuable tools for investigating pairing interactions. To understand the influence of direct base–base interactions in relation to the role of water, pairing free energies between natural nucleobases and fluorinated analogs are estimated by potential of mean force calculations. Compared to pairing of AU and GC, pairing involving fluorinated analogs is unfavorable by 0.5–1.0 kcal mol -1. Decomposing the pairing free energies into enthalpic and entropic contributions reveals fundamental differences for Watson–Crick pairs compared to pairs involving fluorinated analogs. These differences originate from direct base–base interactions and contributions of water. Pairing free energies of fluorinated base analogs with natural bases are less unfavorable by 0.5–1.0 kcal mol -1 compared to non-fluorinated analogs. This is attributed to stabilizing C–F…H–N dipolar interactions and stronger N…H–C hydrogen bonds, demonstrating direct and indirect influences of fluorine. 7-methyl-7H-purine and its 9-deaza analog (Z) have been suggested as members of a new class of non-fluorinated base analogs. Z is found to be the least destabilizing universal base in the context of RNA known to date. This is the first experimental evidence for nitrogen-containing heterocylces as bioisosteres of aromatic rings bearing fluorine atoms.

Das Ziel der vorliegenden Arbeit war, zu analysieren, welche Einflussgrößen sich auf die Kryotransferergebnisse positiv oder negativ auswirken können, um somit Paaren, die über kryokonservierte Zellen verfügen, am effektivsten zu einem gemeinsamen Kind zu verhelfen.
Untersucht wurde, ob das Alter der Patientinnen, die Art der Befruchtung, die Anzahl der übertragenen Embryonen und die Zyklusvorbereitung einen Unterschied auf die Entwicklungs- und Implantationsfähigkeit menschlicher Eizellen sowie auf die Schwangerschaftsrate nach Kryokonservierung und Auftauen im Pronucleus-Stadium aufwiesen.
In der vorliegenden Studie wurden 126 Patientinnen mit ungewollter Kinderlosigkeit, die zwischen 2002 und 2004 am Universitätsklinikum Frankfurt am Main in Behandlung waren, aufgenommen. Das Durchschnittsalter der Patientinnen lag bei 33,3 Jahren. Von 226 durchgeführten Kryotransferzyklen resultierten die Pronukleus-Eizellen bei 34 Kryotransferzyklen aus einer In-vitro-Fertilisation (IVF) und bei 192 Zyklen aus einer intrazytoplasmatischen Spermieninjektion (ICSI).
Es wurde festgestellt, dass in der vorliegenden Studie die erzielte Erfolgsrate, gemessen an der relativ geringen Schwangerschaftsrate, deutlich niedriger lag als beim deutschen IVF-Register. Das Resultat muss jedoch auch unter dem Aspekt der schlechten Ergebnisse der IVF-/ICSI-Therapie in der untersuchten Gruppe betrachtet werden.
Die Ergebnisse der Kryotherapie sind dagegen mit den Zahlen anderer Reproduktionszentren vergleichbar, die derzeit eine Effektivität von durchschnittlich 19,12 % erreichen (DIR 2007) (23).
Bei der Untersuchung wurde jeweils nur der erste und zweite Kryotransferzyklus pro Patientin ausgewertet, damit die Voraussetzung zur Untersuchung der Einflüsse anamnestischer Faktoren gleich war.
Durch ICSI befruchtete Eizellen führten nach Kryokonservierung seltener zu einer Schwangerschaft (11,59%) als nach IVF (16,66%), beim Primärtransfer war das Ergebnis weniger verschieden (9,41% und 7,40%).
Die Vorbereitung des Kryotransferzylus war insofern relevant, als das der Transfer im spontanen Zyklus bezüglich der Schwangerschaftsrate am erfolgreichsten war (13,79%). Der Transfer in einem artifiziellen Zyklus, also nach Stimulation aus einer Kombination von HMG mit FSH, der alleinigen Verabreichung beider Medikamente oder nach dem Kaufmann-Schema, erbrachte dagegen nur eine geringe Schwangerschaftsrate von 6,45%.
Signifikante Unterschiede wiesen die im Punktionszyklus verwendeten Medikamente zur ovariellen Stimulation hinsichtlich der Degenerationsrate nach dem Auftauen der PN-Stadien auf, die nach HMG-Stimulation niedriger (19,35%) als nach FSH-Stimulation (30,82%) war, die Schwangerschaftsrate wies allerdings keine deutlichen Unterschiede auf (11,90% und 13,08%).
Das Alter der Patientinnen hatte wider Erwarten in der vorliegenden Untersuchung keinen eindeutigen Einfluss auf die Schwangerschaftsrate.
Die Resultate bestätigten, dass ein Transfer von maximal drei erlaubten Embryonen die besten Ergebnisse erzielte (19,6 %), hingegen bei einem Transfer von zwei Embryonen ein Ergebnis von nur 5,5 % vorlag. Dieses Ergebnis spricht, wie bereits bei anderen Forschungsergebnissen postuliert, für einen Transfer von drei Embryonen und sollte somit angestrebt werden.
Es lässt sich zusammenfassen, dass die Ergebnisse der Kryotransferbehandlungen in Bezug auf die untersuchten Parameter, wie spontan/hormonelle Kryotransferzyklen, IVF- oder ICSI- Behandlung und Alter, nur geringe Unterschiede aufwiesen.
Einen signifikanten Einfluss auf die Erfolgsrate hatten jedoch die Anzahl der transferierten Embryonen und die verschiedenen Stimulationsmedikamente auf die Degeneration der Oozyten.
Die vorliegende Arbeit hat gezeigt, dass die Kryokonservierung die Effizienz der reproduktionsmedizinischen Behandlung nachweislich erhöht. Darüber hinaus plädiert sie für ein Überdenken des deutschen Embryonenschutzgesetzes, um somit den Frauen eine hohe Anzahl von Behandlungszyklen zu ersparen.
Rhinoliths are mineralised foreign bodies in the nasal cavity that are a chance finding at anterior rhinoscopy. Undiscovered, they grow appreciably in size and can cause a foul-smelling nasal discharge and breathing problems. Giant nasal stones are now a very rare occurrence, since improved diagnostic techniques, such as endoscopic/microscopic rhinoscopy, now make it possible to identify foreign bodies at an early stage of development. We report the case of a 37-year-old patient who, at the age of 5-6 years, introduced a foreign body, probably a stone, into his right nasal cavity. On presentation, he complained of difficulty in breathing through the right nostril that had persisted for the last 10 years. For the past four years a strong fetid smell from the nose had been apparent to those in his vicinity. Under general anaesthesia, the stone was removed in toto from the right nasal cavity. The possible genesis of the rhinolith is discussed, our case compared with those described in the literature, and possible differential diagnoses are considered.
Peroxisome proliferator-activated receptor γ (PPARγ) gained considerable interest as a therapeutic target during chronic inflammatory diseases. Remarkably, the pathogenesis of diseases such as multiple sclerosis or Alzheimer is associated with impaired PPARγ expression. Considering that regulation of PPARγ expression during inflammation is largely unknown, we were interested in elucidating underlying mechanisms. To this end, we initiated an inflammatory response by exposing primary human macrophages to lipopolysaccharide (LPS) and observed a rapid decline of PPARγ1 expression. Because promoter activities were not affected by LPS, we focused on mRNA stability and noticed a decreased mRNA half-life. As RNA stability is often regulated via 3′-untranslated regions (UTRs), we analyzed the impact of the PPARγ-3′-UTR by reporter assays using specific constructs. LPS significantly reduced luciferase activity of the pGL3-PPARγ-3′-UTR, suggesting that PPARγ1 mRNA is destabilized. Deletion or mutation of a potential microRNA-27a/b (miR-27a/b) binding site within the 3′-UTR restored luciferase activity. Moreover, inhibition of miR-27b, which was induced upon LPS exposure, partially reversed PPARγ1 mRNA decay, whereas miR-27b overexpression decreased PPARγ1 mRNA content. In addition, LPS further reduced this decay. The functional relevance of miR-27b-dependent PPARγ1 decrease was proven by inhibition or overexpression of miR-27b, which affected LPS-induced expression of the pro-inflammatory cytokines tumor necrosis factor α (TNFα) and interleukin (IL)-6. We provide evidence that LPS-induced miR-27b contributes to destabilization of PPARγ1 mRNA. Understanding molecular mechanisms decreasing PPARγ might help to better appreciate inflammatory diseases.
The human immunodeficiency virus (HIV) protease inhibitor saquinavir shows anticancer activity. Although its nitric oxide-modified derivative saquinavir-NO (saq-NO) was less toxic to normal cells, it exerted stronger inhibition of B16 melanoma growth in syngeneic C57BL/6 mice than saquinavir did. Saq-NO has been shown to block proliferation, upregulate p53 expression, and promote differentiation of C6 glioma and B16 cells. The anticancer activity of substances is frequently hampered by cancer cell chemoresistance mechanisms. Therefore, we here investigated the roles of p53 and the ATP-binding cassette (ABC) transporters P-glycoprotein (P-gp), multidrug resistance-associated protein 1 (MRP1), and breast cancer resistance protein 1 (BCRP1) in cancer cell sensitivity to saq-NO to get more information about the potential of saq-NO as anticancer drug. Saq-NO exerted anticancer effects in lower concentrations than saquinavir in a panel of human cancer cell lines. Neither p53 mutation or depletion nor expression of P-gp, MRP1, or BCRP1 affected anticancer activity of saq-NO or saquinavir. Moreover, saq-NO sensitized P-gp-, MRP1-, or BCRP1-expressing cancer cells to chemotherapy. Saq-NO induced enhanced sensitization of P-gp- or MRP1-expressing cancer cells to chemotherapy compared with saquinavir, whereas both substances similarly sensitized BCRP1-expressing cells. Washout kinetics and ABC transporter ATPase activities demonstrated that saq-NO is a substrate of P-gp as well as of MRP1. These data support the further investigation of saq-NO as an anticancer drug, especially in multidrug-resistant tumors.
Transcripts of NANOG and OCT4 have been recently identified in human t(4;11) leukemia and in a model system expressing both t(4;11) fusion proteins. Moreover, downstream target genes of NANOG/OCT4/SOX2 were shown to be transcriptionally activated. However, the NANOG1 gene belongs to a gene family, including a gene tandem duplication (named NANOG2 or NANOGP1) and several pseudogenes (NANOGP2-P11). Thus, it was unclear which of the NANOG family members were transcribed in t(4;11) leukemia cells. 5'-RACE experiments revealed novel 5'-exons of NANOG1 and NANOG2, which could give rise to the expression of two different NANOG1 and three different NANOG2 protein variants. Moreover, a novel PCR-based method was established that allows distinguishing between transcripts deriving from NANOG1, NANOG2 and all other NANOG pseudogenes (P2–P11). By applying this method, we were able to demonstrate that human hematopoietic stem cells and different leukemic cells transcribe NANOG2. Furthermore, we functionally tested NANOG1 and NANOG2 protein variants by recombinant expression in 293 cells. These studies revealed that NANOG1 and NANOG2 protein variants are functionally equivalent and activate a regulatory circuit that activates specific stem cell genes. Therefore, we pose the hypothesis that the transcriptional activation of NANOG2 represents a ‘gain-of-stem cell function’ in acute leukemia.
Epoxyeicotrienoic acids (EETs) are cytochrome P450-dependent anti-hypertensive and anti-inflammatory derivatives of arachidonic acid, which are highly abundant in the kidney and considered reno-protective. EETs are degraded by the enzyme soluble epoxide hydrolase (sEH) and sEH inhibitors are considered treatment for chronic renal failure (CRF). We determined whether sEH inhibition attenuates the progression of CRF in the 5/6-nephrectomy model (5/6-Nx) in mice. 5/6-Nx mice were treated with a placebo, an ACE-inhibitor (Ramipril, 40 mg/kg), the sEH-inhibitor cAUCB or the CYP-inhibitor fenbendazole for 8 weeks. 5/6-Nx induced hypertension, albuminuria, glomerulosclerosis and tubulo-interstitial damage and these effects were attenuated by Ramipril. In contrast, cAUCB failed to lower the blood pressure and albuminuria was more severe as compared to placebo. Plasma EET-levels were doubled in 5/6 Nx-mice as compared to sham mice receiving placebo. Renal sEH expression was attenuated in 5/6-Nx mice but cAUCB in these animals still further increased the EET-level. cAUCB also increased 5-HETE and 15-HETE, which derive from peroxidation or lipoxygenases. Similar to cAUCB, CYP450 inhibition increased HETEs and promoted albuminuria. Thus, sEH-inhibition failed to elicit protective effects in the 5/6-Nx model and showed a tendency to aggravate the disease. These effects might be consequence of a shift of arachidonic acid metabolism into the lipoxygenase pathway.
Background: A delta and C fibers are the major pain-conducting nerve fibers, activate only partly the same brain areas, and are differently involved in pain syndromes. Whether a stimulus excites predominantly A delta or C fibers is a commonly asked question in basic pain research but a quick test was lacking so far. Methodology/Principal Findings: Of 77 verbal descriptors of pain sensations, "pricking", "dull" and "pressing" distinguished best (95% cases correctly) between A delta fiber mediated (punctate pressure produced by means of von Frey hairs) and C fiber mediated (blunt pressure) pain, applied to healthy volunteers in experiment 1. The sensation was assigned to A delta fibers when "pricking" but neither "dull" nor "pressing" were chosen, and to C fibers when the sum of the selections of "dull" or "pressing" was greater than that of the selection of "pricking". In experiment 2, with an independent cohort, the three-descriptor questionnaire achieved sensitivity and specificity above 0.95 for distinguishing fiber preferential non-mechanical induced pain (laser heat, exciting A delta fibers, and 5-Hz electric stimulation, exciting C fibers). Conclusion: A three-item verbal rating test using the words "pricking", "dull", and "pressing" may provide sufficient information to characterize a pain sensation evoked by a physical stimulus as transmitted via A delta or via C fibers. It meets the criteria of a screening test by being easy to administer, taking little time, being comfortable in handling, and inexpensive while providing high specificity for relevant information.
Leukotrienes constitute a group of bioactive lipids generated by the 5-lipoxygenase (5-LO) pathway. An increasing body of evidence supports an acute role for 5-LO products already during the earliest stages of pancreatic, prostate, and colorectal carcinogenesis. Several pieces of experimental data form the basis for this hypothesis and suggest a correlation between 5-LO expression and tumor cell viability. First, several independent studies documented an overexpression of 5-LO in primary tumor cells as well as in established cancer cell lines. Second, addition of 5-LO products to cultured tumor cells also led to increased cell proliferation and activation of anti-apoptotic signaling pathways. 5-LO antisense technology approaches demonstrated impaired tumor cell growth due to reduction of 5-LO expression. Lastly, pharmacological inhibition of 5-LO potently suppressed tumor cell growth by inducing cell cycle arrest and triggering cell death via the intrinsic apoptotic pathway. However, the documented strong cytotoxic off-target effects of 5-LO inhibitors, in combination with the relatively high concentrations of 5-LO products needed to achieve mitogenic effects in cell culture assays, raise concern over the assignment of the cause, and question the relationship between 5-LO products and tumorigenesis. Background: Descending inhibitory pain control contributes to the endogenous defense against chronic pain and involves noradrenergic and serotonergic systems. The clinical efficacy of antidepressants suggests that serotonin may be particularly relevant for neuropathic pain conditions. Serotonergic signaling is regulated by synthesis, metabolisms, reuptake and receptors. To address the complexity, we used inbred mouse strains, C57BL/6J, 129 Sv, DBA/2J and Balb/c, which differ in brain serotonin levels. Results: Serotonin analysis after nerve injury revealed inter-strain differences in the adaptation of descending serotonergic fibers. Upregulation of spinal cord and midbrain serotonin was apparent only in 129 Sv mice and was associated with attenuated nerve injury evoked hyperalgesia and allodynia in this strain. The increase of dorsal horn serotonin was blocked by hemisectioning of descending fibers but not by rhizotomy of primary afferents indicating a midbrain source. Para-chlorophenylalanine-mediated serotonin depletion in spinal cord and midbrain intensified pain hypersensitivity in the nerve injury model. In contrast, chronic inflammation of the hindpaw did not evoke equivalent changes in serotonin levels in the spinal cord and midbrain and nociceptive thresholds dropped in a parallel manner in all strains. Conclusion: The results suggest that chronic nerve injury evoked hypernociception may be contributed by genetic differences of descending serotonergic inhibitory control.
Background: R-flurbiprofen, one of the enantiomers of flurbiprofen racemate, is inactive with respect to cyclooxygenase inhibition, but shows analgesic properties without relevant toxicity. Its mode of action is still unclear. Methodology/Principal Findings: We show that R-flurbiprofen reduces glutamate release in the dorsal horn of the spinal cord evoked by sciatic nerve injury and thereby alleviates pain in sciatic nerve injury models of neuropathic pain in rats and mice. This is mediated by restoring the balance of endocannabinoids (eCB), which is disturbed following peripheral nerve injury in the DRGs, spinal cord and forebrain. The imbalance results from transcriptional adaptations of fatty acid amide hydrolase (FAAH) and NAPE-phospholipase D, i.e. the major enzymes involved in anandamide metabolism and synthesis, respectively. R-flurbiprofen inhibits FAAH activity and normalizes NAPE-PLD expression. As a consequence, R-Flurbiprofen improves endogenous cannabinoid mediated effects, indicated by the reduction of glutamate release, increased activity of the anti-inflammatory transcription factor PPAR gamma and attenuation of microglia activation. Antinociceptive effects are lost by combined inhibition of CB1 and CB2 receptors and partially abolished in CB1 receptor deficient mice. R-flurbiprofen does however not cause changes of core body temperature which is a typical indicator of central effects of cannabinoid-1 receptor agonists. Conclusion: Our results suggest that R-flurbiprofen improves the endogenous mechanisms to regain stability after axonal injury and to fend off chronic neuropathic pain by modulating the endocannabinoid system and thus constitutes an attractive, novel therapeutic agent in the treatment of chronic, intractable pain.
I-kappaB-Kinase epsilon - ein neues Zielprotein für die Pharmakotherapie bei Schmerz und Entzündung?

Der Transkriptionsfaktor NF-kappaB spielt eine wichtige Rolle bei der Regulation von Immunantworten, Apoptose und Entzündungen sowie bei der Entstehung und Verarbeitung von Schmerzen. Ein pharmakologischer Eingriff in die NF-kappaB-Aktivierungskaskade könnte daher eine Schmerzhemmung bewirken und so Ansätze für die Entwicklung neuer Therapien für pathophysiologische Schmerzen liefern. Die NF-kappaB-Signalübertragungskaskade bietet verschiedene Angriffspunkte für Pharmaka, wobei zurzeit IkappaB Kinasen (IKK) als hoffnungsvolle Zielmoleküle im Fokus der Untersuchungen stehen. Verschiedene IKKs regulieren die Aktivität von NF-kappaB über die Phosphorylierung des inhibitorischen Proteins IkappaB oder über die direkte Phosphorylierung von NF-kappaB. In der vorliegenden Arbeit wurde die Rolle der neu entdeckten IKK epsilon bei der Schmerzentstehung und -verarbeitung sowie deren Eignung als neues Zielmolekül für die Schmerztherapie näher untersucht. Es konnte gezeigt werden, dass IKK epsilon konstitutiv in Geweben der Maus, welche an der Entstehung und Verarbeitung von Schmerzen beteiligt sind, exprimiert ist. Im Rückenmark konnte die Lokalisation von IKK epsilon in den schmerzrelevanten Laminae I und II des Dorsalhorns nachgewiesen werden und auch in den Hinterwurzelganglien (Dorsal Root Ganglia (DRG’s)) war IKK epsilon in kleinen, nozizeptiven Neuronen exprimiert. Nach peripherer entzündlich-nozizeptiver Stimulation mit Formalin oder Zymosan kam es im Lumbalmark und den DRG’s zu einem signifikanten Anstieg der IKK epsilon-Expression sowohl auf mRNA- als auch auf Proteinebene. Diese Beobachtungen machten eine Beteiligung von IKK epsilon an der Prozessierung von Schmerz sehr wahrscheinlich. Um die Rolle von IKK epsilon während der Schmerzentstehung und -verarbeitung besser beurteilen zu können wurde das Verhalten von IKK epsilon defizienten Mäusen in akuten und inflammatorischen Schmerzmodellen charakterisiert. Es konnte gezeigt werden, dass der Knockout von IKK epsilon zu einem signifikant verringerten nozizeptiven Verhalten im Formalintest und einer Hemmung der mechanischen Hyperalgesie nach Zymosaninjektion im Vergleich zu Wildtyp-Mäusen führte. Gleichzeitig konnte kein Unterschied im akut nozizeptiven Verhalten festgestellt werden. Der Knockout von IKK epsilon hatte demnach keine Auswirkung auf den akuten physiologischen Nozizeptorschmerz, zeigte jedoch eine Verbesserung bei pathophysiologischen Schmerzen. Das verringerte nozizeptive Verhalten der IKK epsilon defizienten Mäuse im Formalintest ging mit einer Hemmung der NF-kappaB-Aktivierung im Rückenmark einher. Auch konnte eine verringerte mRNA-Expression der NF-kappaB-abhängigen Gene Cyclooxygenase-2 (COX-2), Matrixmetalloprotease-9 (MMP-9) und induzierbare Stickstoffmonoxid-Synthase (iNOS), die an der Regulation von Entzündungsschmerzen beteiligt sind, im Rückenmark und den DRG’s nachgewiesen werden. Da IKK epsilon bisher hauptsächlich mit der Aktivierung des TypI Interferon-Signalweges in Zusammenhang gebracht wurde, wurde außerdem geprüft, ob es nach Injektion mit Formalin zu einer Aktivierung des Trankriptionsfaktors Interferon-regulierender Faktor (IRF)-3 in Wildtyp-Mäusen kommt, was nicht beobachtet werden konnte. Der Knockout von IKK epsilon scheint demnach seine antinozizeptive Wirkung direkt über eine fehlende Aktivierung von NF-kappaB zu entfalten, wonach IKK epsilon eine bedeutendere Rolle als bisher angenommen bei der Aktivierung von NF-kappaB spielt. Dies konnte durch in vitro Daten untermauert werden. Der Knockdown von IKK epsilon in Makrophagen-Zellkultur mit spezifischer siRNA verhinderte die Phosphorylierung von NF-kappaBp65 am Serinrest 536 nach Stimulation mit LPS. Anhand der vorliegenden Daten lässt sich also schlussfolgern, dass IKK epsilon an der Schmerzentstehung und verarbeitung bei Entzündungen beteiligt zu sein scheint. Eine Hemmung dieser Kinase könnte demnach ein neues, lohnendes Ziel für die Entwicklung neuer Medikamente für die Schmerztherapie sein.
In the past, the genetically diabetic-obese diabetes/diabetes (db/db) and obese/obese (ob/ob) mouse strains were used to investigate mechanisms of diabetes-impaired wound healing. Here we determined patterns of skin repair in genetically normal C57Bl/6J mice that were fed using a high fat diet (HFD) to induce a diabetes-obesity syndrome. Wound closure was markedly delayed in HFD-fed mice compared to mice which had received a standard chow diet (CD). Impaired wound tissue of HFD mice showed a marked prolongation of wound inflammation. Expression of vascular endothelial growth factor (VEGF) was delayed and associated with the disturbed formation of wound margin epithelia and an impaired angiogenesis in the reduced granulation tissue. Normal wound contraction was retarded and disordered. Wound disorders in obese C57Bl/6J mice were paralleled by a prominent degradation of the inhibitor of NFκB (IκB-α) in the absence of an Akt activation. By contrast to impaired wound conditions in ob/ob mice, late wounds of HFD mice did not develop a chronic inflammatory state and were epithelialized after 11 days of repair. Thus, only genetically obese and diabetic ob/ob mice finally developed chronic wounds and therefore represent a better suited experimental model to investigate diabetes-induced wound healing disorders.
Die Wirkungen von Acrylamid beschäftigen seit Jahrzehnten Mediziner, Toxikologen, Biologen und Analytiker. Zwar konnte eine kanzerogene Wirkung beim Tier eindeutig nachgewiesen werden, der Nachweis dieser Wirkung beim Menschen ist aber bis heute nicht erbracht. Bis 2000 wurde angenommen, dass Acrylamid ausschließlich technisch erzeugt vorkommt und der Mensch Acrylamid nur aus dieser Quelle aufnimmt. Hierauf beruhen Grenz- und Richtwerte auf nationaler und internationaler Ebene, sowohl am Arbeitsplatz als auch für Trinkwasser und Verpackungsmaterialien. Neben dem Acrylamidgehalt in technischen Produkten, Wasser und Luft gelang der Nachweis der aufgenommenen Acrylamidmenge über Acrylamid-Hämoglobin-Addukte im Blut erst 1995, nachdem der Acrylamid-Metabolismus aufgeklärt worden war und die analytischen Methoden verfeinert waren. Die Annahme, dass Acrylamid ausschließlich in technischen Prozessen gebildet wird, war jedoch nicht vereinbar mit Erkenntnissen, dass auch Probanden einen erhöhten Acrylamidspiegel im Blut aufzeigten, die nicht mit Acrylamid am Arbeitsplatz in Kontakt gekommen waren. Im Jahr 2002 gelang es eindeutig nachzuweisen, dass Acrylamid in vielen, meist alltäglich verzehrten Lebensmitteln abhängig von der Zubereitungsart zu finden ist. Mit Hilfe einer verbesserten Analytik konnten schließlich hohe Acrylamidmengen in zahlreichen, vor allem frittierten Lebensmitteln, belegt werden, die die Belastung aus technischen Quellen oft weit überschritten. Die Tatsache, dass die Nahrung der allgemeinen Bevölkerung deutlich Acrylamid-belastet ist, hat die Bewertung der Acrylamid-Toxizität und die Maßnahmen zum Gesundheitsschutz nachhaltig verändert. Durch den späten Nachweis der Acrylamid-Aufnahme mit Lebensmitteln sind viele frühere Annahmen für die Gesundheitsbewertung irrig, die hieraus gezogenen Schlußfolgerungen nicht haltbar. Seit 2002 ist die Reduktion der Lebensmittelbelastung in den Mittelpunkt gerückt, die Arbeitsplatzbelastung hat - mit Ausnahme der Schleimhautreizung - keine weitere Berücksichtigung erhalten. Die hohe Belastung von Lebensmitteln stellt bisherige Richtlinien und Expositionsauflagen in Frage, der fehlende bisherige epidemiologische Nachweis, der aus Tierversuchen eindeutig nachweisbaren Karzinogenität stellt für Acrylamid auch die bisherigen Modell-Annahmen der Krebsentstehung durch Umweltschadstoffe in der „one hit-Hypothese“ in Frage.
This paper introduces a new methodology for the fabrication of strain-sensor elements for MEMS and NEMS applications based on the tunneling effect in nano-granular metals. The strain-sensor elements are prepared by the maskless lithography technique of focused electron-beam-induced deposition (FEBID) employing the precursor trimethylmethylcyclopentadienyl platinum [MeCpPt(Me)3]. We use a cantilever-based deflection technique to determine the sensitivity (gauge factor) of the sensor element. We find that its sensitivity depends on the electrical conductivity and can be continuously tuned, either by the thickness of the deposit or by electron-beam irradiation leading to a distinct maximum in the sensitivity. This maximum finds a theoretical rationale in recent advances in the understanding of electronic charge transport in nano-granular metals.
Die Genexpression in prokaryotischen Organismen unterliegt einer Vielzahl von Regulationsmechanismen, deren Aufgabe darin besteht, die Zelle an sich ändernde Umweltbedingungen anzupassen, um so das Überleben des prokaryotischen Organismus zu gewährleisten. Eine Reihe von Hitzeschock- und Virulenzgenen unterliegen temperaturabhängiger Regulation, mit dem Ziel, die Zelle an die sich ändernde Umgebung anzupassen. Die Messung der Temperatur erfolgt dabei über temperatursensitive RNA-Elemente, sogenannte RNA-Thermometer, die sich üblicherweise in der 5’-untranslatierten Region der Gene befinden, die sie regulieren. Sie unterdrücken die Translationsinitiation, indem sie die Shine-Dalgarno (SD)-Sequenz bei niedrigen Temperaturen über Basenpaarung blockieren und dadurch die Bindung des Ribosoms verhindern. In Kapitel 2 der vorliegenden Arbeit wurde die thermodynamische Stabilität der temperatursensitiven Haarnadelschleife 2 des Salmonella FourU RNA-Thermometers über einen breiten Temperaturbereich analysiert. Freie Enthalpie-, Enthalpie- und Entropie-Werte für die Basenpaaröffnung der einzelnen Nukleobasen innerhalb der RNA wurden über die temperaturabhängige Messung von Iminoprotonen-Austauschraten mittels NMR-Spektroskopie bestimmt. Die Austauschraten wurden für die Wildtyp-RNA und die A8C-Mutante bestimmt und miteinander verglichen. Es zeigte sich, dass die Wildtyp-RNA durch das außergewöhnlich stabile Basenpaar G14-C25 stabilisiert wird. Dies konnte durch die Untersuchung der Entfaltung der destabilisierenden G14A-C25U-Doppelmutante verifiziert werden. Über CD-spektroskopsiche Untersuchungen konnte der globale Entfaltungsübergang der jeweiligen RNA analysiert werden. Das Mismatch-Basenpaar innerhalb des Wildtyp-RNA-Thermometers (A8-G31) erwies sich als Ursache für die geringere Kooperativität des Entfaltungsübergangs der Wildtyp-RNA im Vergleich zur A8C-Mutante. Enthalpie- und Entropie-Werte für die Basenpaaröffnung einzelner Nukleotide sind für beide RNAs linear korreliert. Die Steigungen dieser Korrelationen stimmen mit den Schmelzpunkten der RNAs überein, die über CD-Spektroskopie bestimmt wurden. Entfaltung der RNA tritt also genau dann auf, wenn alle Nukleotide gleiche thermodynamische Stabilitäten besitzen. Die Resultate sind mit einem Reißverschluss-Mechanismus für die RNA-Helix Entfaltung konsistent und erklärbar, in dem die Stapelinteraktionen der benachbarten Nukleobasen innerhalb der RNA-Helix verantwortlich für die beobachtete Kooperativität sind. Die Ergebnisse weisen auch auf die Wichtigkeit der RNA-Lösungsmittel-Interaktion für die Stabilität der RNA-Struktur hin. So konnten langreichweitige Wechselwirkungen der A8C-Mutation auf die Stabilität der G14-Nukleobase identifiziert werden, die möglicherweise über die Hydrathülle der RNA vermittelt werden. Schließlich konnte für das FourU-Motiv eine Mg2+-Bindestelle identifiziert werden, die die temperaturabhängige Stabilität des RNA-Thermometers beeinflusst. Es besteht also die Möglichkeit, dass Änderungen der intrazellulären Mg2+-Konzentration die Expression des agsA-Gens in vivo modulierend beeinflussen. In Kapitel 3 dieser Arbeit wurden die dynamischen Eigenschaften des Phosphodiesterrückgrats einer perdeuterierten cUUCGg-Tetraloop-14mer-RNA untersucht. Dazu wurden die Relaxationseigenschaften aller 31P-Kerne dieser RNA bei magnetischen Feldstärken von 300, 600 und 900 MHz untersucht. Dipolare Relaxationsbeiträge konnten unterdrückt werden, indem eine perdeuterierte RNA-Probe in einem D2O-Puffer verwendet wurde. Um die 31P-Relaxationsdaten (R1, R2) interpretieren zu können, wurde zusätzlich mittels Festkörper-NMR die Chemische Verschiebungsanisotropie (CSA) der 31P-Kerne des Phosphodiesterrückgrats bestimmt. Die Messungen wurden bei verschiedenen Salzkonzentrationen und unter unterschiedlichen Hydratationsbedingungen durchgeführt. Aus den Daten konnte ein 31P-CSA-Wert von 178.5 ppm im statischen Zustand (S2 = 1) bestimmt werden. Auf der Grundlage der durchgeführten R1- und R2-Messungen wurde eine Modelfree-Analyse durchgeführt, um Informationen über die schnellen Dynamiken des Phosphodiesterrückgrats zu erhalten. Die Resultate zeigen, dass die Dynamiken des Phosphodiesterrückgrats auf der Subnanosekundenzeitskala stärker ausgeprägt sind als die Dynamiken der Ribofuranosylreste und der Nukleobasen. Des Weiteren konnte gezeigt werden, dass die Dynamik einer individuellen Phosphatgruppe zu der jeweiligen 5’-benachbarten Nukleobase korreliert ist. In Kapitel 4 dieser Arbeit wird die Entwicklung neuer Methoden beschrieben, mit denen Torsionswinkelinformation aus der Analyse kreuzkorrelierter Relaxationsraten gewonnen werden können. Im ersten Teil des Kapitels wird die Entwicklung einer neuen NMR-Pulssequenz beschrieben, über die der glykosidische Torsionswinkel Chi in 13C,15N-markierten Oligonukleotiden bestimmt werden kann. Mit dem neuen quantitativen Gamma-HCNCH-Experiment ist es möglich, die dipolaren kreuzkorrelierten Relaxationsraten Gamma-C6H6-C1´H1´ (Pyrimidine) und Gamma-C6H6-C1´H1´ (Purine) zu messen. Die kreuzkorrelierten Relaxationsraten wurden an einer 13C,15N-markierten cUUCGg-Tetraloop-14mer-RNA bestimmt. Die aus den Raten extrahierten Chi-Winkel wurden mit bereits vorhandener Strukturinformation verglichen. Sie stimmen bemerkenswert gut mit den Winkeln der Kristallstruktur des Tetraloops überein. Zusätzlich wurde die neue Methode an einer größeren 30mer-RNA, dem „Stemloop D“ (SLD) aus dem Coxsackievirus-B3-Kleeblatt, getestet. Für die SLD-RNA wurde der Effekt von anisotroper Rotationsdiffusion auf die Relaxationsraten untersucht. Es konnte gezeigt werden, dass die Chi-Winkelbestimmung besonders für Nukleotide in der anti-Konformation sehr genau ist und die Methode eine eindeutige Unterscheidung von syn- und anti-Konformation zulässt. Im zweiten Teil von Kapitel 4 wird die Entwicklung des Gamma-HCCCH-Experiments beschrieben. Hierbei handelt es sich um eine neue NMR-Pulssequenz zur Messung der Gamma-C1´H1´-C3´H3´-Rate in 13C-markierten RNAs. Die Funktionsfähigkeit der neuen Methode wurde an einer cUUCGg-Tetraloop-14mer-RNA demonstriert. Zusätzlich dazu wurden die analytischen Gamma-C1´H1´-C3´H3´(P,nü_max)-, Gamma-C1´H1´-C4´H4´(P,nü_max)- und Gamma-C2´H2´-C4´H4´(P,nü_max)-Abhängigkeiten mathematisch hergeleitet. Die an der 14mer-RNA gemessenen Gamma-C1´H1´-C3´H3´-Raten wurden mit Hilfe der Gamma-C1´H1´-C3´H3´(P,nü_max)-Beziehung analysiert. Die Ergebnisse für die Pseudorotationsphase P sind konsistent mit Referenzwinkeln aus der 14mer-NMR-Struktur und den bereits bekannten (Gamma-C1´H1´-C2´H2´)/(Gamma-C3´H3´-C4´H4´)-Ratenverhältnissen. Die neue Methode liefert zusätzliche Informationen, um Konformation (P, nü_max) und Dynamik S2(C1´H1´-C3´H3´) der Ribosereste in RNA-Molekülen genauer beschreiben zu können. In Kapitel 5 dieser Arbeit wird die Entwicklung des 3D-HNHC-Experiments, einer neuen NMR-Pulssequenz, beschrieben. Dieses Experiment ermöglicht es, die H2-, C2- und N1-Resonanzen in Adenin-Nukleobasen 13C, 15N-markierter RNA-Oligonukleotide miteinander zu korrelieren. Die Funktionsfähigkeit der neuen Methode wurde an einer mittelgroßen, entsprechend markierten 36mer-RNA demonstriert. Die neue Methode vereinfacht die Zuordnung der Kerne der Adenin-Nukleobasen, da Zuordnungsmehrdeutigkeiten aufgrund überlappender Resonanzen in der 1H-Dimension aufgelöst werden können. In Kombination mit dem TROSY-relayed-HCCH-COSY-Experiment liefert das neue 3D-HNHC-Experiment das fehlende Glied für die Zuordnung der Imino-H3-Resonanzen der Uracil-Nukleobasen über das AU-Basenpaar hinweg zu den H8-Resonanzen der Adenin-Nukleobasen.
Die Arbeit untersucht die Herstellung und ökonomische Umsetzung von "ethnischem Anderssein" in sog. Kultur-Kochkursen in Frankfurt am Main. In den beforschten Kultur-Kochkursen vermitteln ethnisch markierte KochkursleiterInnen "ihre Küche und Kultur" einem (multikulturalistisch) interessiertem Publikum. Die Arbeit fokussiert auf die performative Herstellung, Umsetzung und Repräsentation von (vermarktbarem) ethnischen Andersseins durch die KochkursleiterInnen in der Kochkurssituation und überprüft die Annahme, dass Ethnizität primär durch (Selbst-)Othering – die Betonung von unüberbrückbarer, essentieller kultureller Differenz – vermarktet wird. Die Befunde, welche auf Internetrecherchen, teilnehmenden Beobachtungen und narrativen Interviews basieren, zeigen, dass die organisierenden Institutionen die Kulturkochkurse zwar mit dem Verweis auf Authentizität, Tradition und Andersartigkeit bewerben, die Kultur-KochkursleiterInnen selbst in der Kochkurssituation allerdings kein kulturalistisches, exotisierendes und Differenz betonendes Ethnizitätskonzept perfomieren, sondern vielmehr Dynamik, Vielfalt und Vermischungen artikulieren, praktizieren und stark positiv konnotieren.
Tubular carbonate concretions of up to 1 m in length and perpendicular to bedding, occur abundantly in the Upper Pliensbachian (upper Amaltheus margaritatus Zone, Gibbosus Subzone) in outcrops (Fontaneilles section) in the vicinity of Rivière-sûr-Tarn, southern France. Stable isotope analyses of these concretions show negative δ13C values that decrease from the rim to the center from −18.8‰ to −25.7‰ (V-PDB), but normal marine δ18O values (−1.8‰). Carbon isotope analyses of Late Pliensbachian bulk carbonate (matrix) samples from the Fontaneilles section show clearly decreasing C-isotope values across the A. margaritatus Zone, from +1‰ to −3‰ (V-PDB). Isotope analyses of coeval belemnite rostra do not document such a negative C-isotope trend with values remaining stable around +2‰ (V-PDB). Computer tomographic (CT) scanning of the tubular concretions show multiple canals that are lined or filled entirely with pyrite. Previously, the formation of these concretions with one, two, or more central tubes, has been ascribed to the activity of an enigmatic organism, possibly with annelid or arthropod affinities, known as Tisoa siphonalis. Our results suggest tisoan structures are abiogenic. Based on our geochemical analyses and sedimentological observations we suggest that these concretions formed as a combination of the anaerobic oxidation of methane (AOM) and sulfate reduction within the sediment. Fluids rich in methane and/or hydrocarbons likely altered local bulk rock carbon isotope records, but did not affect the global carbon cycle. Interestingly, Tisoa siphonalis has been described from many locations in the Grands Causses Basin in southern France, and from northern France and Luxemburg, always occurring at the same stratigraphic level. Upper Pliensbachian authigenic carbonates thus possibly cover an area of many thousand square kilometers. Greatly reduced sedimentation rates are needed to explain the stabilization of the sulfate-methane transition zone in the sedimentary column in order for the tubular concretions to form. Late Pliensbachian cooling, reducing run-off, and/or the influx of colder water and more vigorous circulation could be responsible for a halt in sedimentation. At the same time (thermogenic) methane may have destabilized during a major phase of Late Pliensbachian sea level fall. As such Tisoa siphonalis is more than a geological curiosity, and its further study could prove pivotal in understanding Early Jurassic paleoenvironmental change.
LmrA is a member of the ATP Binding Cassette (ABC) transporter family of membrane proteins and a structural and functional homologue of P-glycoprotein1, 2. ABC-transporters share a common architecture of two transmembrane domains and two nucleotide binding domains. The NBDs are highly conserved in this transporter family whereas the TMDs are highly diverse3. The TMDs recognize the substrate and the NBDs bind and hydrolyze ATP and thus contribute the energy for substrate translocation. ABC transporters as a protein family transport a high number of substrates including peptides, nutrients, ions, bile acids, lipids and other lipophilic compounds. LmrA is a multidrug transporter that recognizes a number of hydrophobic substrates including fluorescent dyes and antibiotics1, 4-6. LmrA is a native protein of the gram-positive bacterium Lactococcus lactis. In this thesis, L. lactis was used as a homologous expression host for the preparation of LmrA for a variety of experiments. Wildtype LmrA as well as a number of cysteine mutants were successfully expressed in L. lactis, purified and subsequently characterized by a variety of biochemical assays (Chapter 4). LmrA can be expressed to very high amounts in L. lactis. The purification and reconstitution were optimized for the requirements of solid-state NMR experiments in this thesis. For the first time, an ABC transporter has been reconstituted in synthetic lipids to a ratio of up to 1:150 (mol/mol). LmrA was shown to be active under magic angle spinning conditions with these reconstitution ratios. By taking advantage of the slower ATP hydrolysis by LmrA ΔK388 (lysine deletion in the Walker A motif), a real-time 31P solid-state NMR ATPase assay was established (Chapter 5). This assay allowed, for the first time, the investigation of all phosphor nuclei during the ATP hydrolysis cycle of a membrane protein simultaneously and in real time7. This assay has been successfully adapted to investigate both ATP hydrolysis and substrate phosphorylation of diacylglycerol kinase (together with S. Wollschlag) and ATP hydrolysis at high temperatures of the thermophilic ABC transporter ABC1 from Thermos thermophilus (together with A. Zutz). In the course of this thesis, the gene for LmrA has been cloned into expression vectors suitable for Escherichia coli and the heterologous expression of LmrA was established (Chapter 4). The functionality of the heterologously expressed protein has been investigated and compared to L. lactis LmrA. In these experiments, LmrA was shown to yield a distinct multidrug resistance phenotype in its E. coli host and to show secondary active multidrug transport in the absence of ATP and presence of a proton gradient [Hellmich et al, in prep] (Chapter 4). Previously, it had been shown that LmrA acts as a seconadary active transporter when the NBDs are truncated8. The overexpression in minimal and defined medium and the purification of LmrA from E. coli have been optimized. Isotope labeling for ssNMR has been established and the first multinuclear ssNMR experiments have been carried out on a functional ABC transporter (Chapter 8). ABC transporters couple two cycles: upon ATP binding, the NBDs dimerize, hydrolyze the ATP, subsequently release Pi and ADP and finally dissociate. During this cycle, conformational changes are relayed to the TMDs which utilize the energy from ATP binding and/or hydrolysis to translocate the respective substrate. The prehydrolysis state can be trapped by beryllium fluoride, whereas the post-hydrolysis state of this cycle can be trapped by vanadate9-12. Trapping protocols for these reagents were successfully established for LmrA in this thesis (Chapter 4). This allowed for the investigation of different catalytic states by both ssNMR and EPR. A general 19F labeling protocol for membrane proteins has been established in the course of this thesis and successfully applied to proteorhodopsin (together with N. Pfleger)13 and LmrA (chapter 6). Single cysteine mutants of LmrA that line out the dimer interface have been labeled with a fluorine label for ssNMR. In the apo state, the 19F labeling indicates highly flexible transmembrane domains, a finding that is supported by 13C ssNMR and EPR measurements. The addition of drugs has a different effect on different positions within the LmrA dimer, therefore indicating that different drugs are recognized at a different position within the protein. For P-glycoprotein and LmrA it has been previously shown by biochemical methods that different drug binding sites co-exist. For a 19F label attached at position 314 (LmrA E314C), the spectra showed two distinct peaks with similar populations. This could hint towards a structural asymmetry within the LmrA dimer that might also be reflected in the alternating ATP hydrolysis at the NBDs. E314 has been specifically implicated with drug transport. Thus, structural asymmetry at this position might be functionally relevant for guiding a substrate through the transporter. Structural asymmetry within a homodimeric ABC transporter has also been shown for BtuCD, the E. coli vitamin B12 importer14. In addition, the conserved glutamates in EmrE, a small multidrug resistance protein, were shown to be asymmetric in the drug bound state15. Both, uniformly 13C/15N labeled as well as selectively amino acid type labeled LmrA has been investigated in different conformational states. Interestingly, significant dynamic changes in the b-sheet regions of LmrA (confined to the NBDs) were observed in the pre-hydrolysis (beryllium fluoride) and transition state (vanadate trapped) state. These were interpreted as the transition from a domain in fast conformational exchange in the apo state to one of intermediate exchange in the nucleotide bound state. A significant change in NBD mobility upon nucleotide binding was previously also shown with 2H ssNMR on LmrA16. By EPR it was shown that LmrA in both the vanadate and BeFx trapped states displays a significantly higher rigidity and therefore defined distances, whereas the apo state resembled a “floppy” protein with no preferred distance distribution. This concurs with data obtained from 19F ssNMR with fluorine labeled single-cysteine mutants. Here, in agreement with the EPR data, a higher label (and possibly) protein mobility was observed in the apo state displaying rather broad line widths. Upon trapping with vanadate, the line widths of the majority of fluorine-labeled mutants decreased due to an enhanced protein rigidity and a more homogenous environment of the fluorine labels. A similar observation was made when increasing the temperature that can be explained due to higher protein flexibility at increased temperatures. Solution NMR was employed to investigate the isolated soluble NBD of LmrA (Chapter 9). First 2D and 3D spectra were successfully obtained and could be utilized for a preliminary assignment of a significant fraction of residues. Additionally, binding of ATP and ADP in absence and presence of magnesium was investigated. Finally, the effects of peptides emulating the coupling helices of the full-length transporter on the soluble NBD were investigated. Strikingly, binding of one of these peptides only occurred in the presence of nucleotides (whereas the other showed no binding at all) hinting towards a tightly coupled regulation of the NBD and TMD during the substrate translocation/ATP hydrolysis cycle based on nucleotide binding.
Tubular carbonate concretions of up to 1 m in length and perpendicular to bedding, occur abundantly in the Upper Pliensbachian (upper Amaltheus margaritatus Zone, Gibbosus Subzone) in outcrops (Fontaneilles section) in the vicinity of Rivière-sûr-Tarn, southern France. Stable isotope analyses of these concretions show negative delta 13C values that decrease from the rim to the center from - 18.8‰ to - 25.7‰ (V-PDB), but normal marine delta 18 O values (- 1.8‰). Carbon isotope analyses of Late Pliensbachian bulk carbonate (matrix) samples from the Fontaneilles section show clearly decreasing C-isotope values across the A. margaritatus Zone, from +1‰ to - 3‰ (V-PDB). Isotope analyses of coeval belemnite rostra do not document such a negative C-isotope trend with values remaining stable around +2‰ (V-PDB). Computer tomographic (CT) scanning of the tubular concretions show multiple canals that are lined or filled entirely with pyrite. Previously, the formation of these concretions with one, two, or more central tubes, has been ascribed to the activity of an enigmatic organism, possibly with annelid or arthropod affinities, known asTisoa siphonalis. Our results suggest tisoan structures are abiogenic. Based on our geochemical analyses and sedimentological observations we suggest that these concretions formed as a combination of the anaerobic oxidation of methane (AOM) and sulfate reduction within the sediment. Fluids rich in methane and/or hydrocarbons likely altered local bulk rock carbon isotope records, but did not affect the global carbon cycle. Interestingly, Tisoa siphonalis has been described from many locations in the Grands Causses Basin in southern France, and from northern France and Luxemburg, always occurring at the same stratigraphic level. Upper Pliensbachian authigenic carbonates thus possibly cover an area of many thousand square kilometers. Greatly reduced sedimentation rates are needed to explain the stabilization of the sulfate-methane transition zone in the sedimentary column in order for the tubular concretions to form. Late Pliensbachian cooling, reducing run-off, and/or the influx of colder water and more vigorous circulation could be responsible for a halt in sedimentation. At the same time (thermogenic) methane may have destabilized during a major phase of Late Pliensbachian sea level fall. As such Tisoa siphonalis is more than a geological curiosity, and its further study could prove pivotal in understanding Early Jurassic paleoenvironmental change.
"Protein Associated with Myc" (PAM) hat aufgrund seiner enormen Größe von 510 kD und der Vielzahl an Proteinbindungsstellen das Potential viele regulatorische und physiologische Prozesse zu regulieren. Mit der Funktion als E3-Ubiquitinligase und davon unabhängigen Proteininteraktionen reguliert es beispielsweise Prozesse der Synaptogenese und der spinalen Schmerzverarbeitung, wie auch die Regulation des Pteridin Stoffwechsels und des cAMP-Signalweges. Im Gegensatz zur spinalen Schmerzverarbeitung war eine Beteiligung von PAM an der peripheren Nozizeption bisher unbekannt und sollte im Rahmen dieser Arbeit untersucht werden. Dazu wurden konditionale PAM-Knockout Mäuse generiert und charakterisiert. Zum einen wurde PAM in Vorläuferzellen von Neuronen und Gliazellen und zum anderen in allen nozizeptiven und thermorezeptiven Neuronen der Dorsalganglia und der Ganglia trigeminale deletiert. Der Knockout in Neuronen und Gliazellen führte zu einer pränatalen Letalität und unterstreicht so die Bedeutung von PAM während der Neuronenentwicklung. Mit Hilfe des spezifischen Knockouts in nozizeptiven Neuronen konnte eine Rolle von PAM bei der Regulation der thermischen Hyperalgesie gezeigt werden. Keinen Einfluss hatte die Deletion von PAM auf basale thermische und mechanische Schmerzschwellen, sowie auf Formalin-induzierte akute Schmerzen. Als potentieller Mechanismus wurde der mTOR- und der p38 MAPK-Signalweg untersucht. Dabei konnte eine Vermittlung der S1P-induzierten mTOR-Aktivierung durch PAM nachgewiesen werden und Rheb als eine Komponente dieser Aktivierung ermittelt werden. Der p38 MAPK Signalweg war in Abwesenheit von PAM konstitutiv aktiviert und einige Proteine des Rezeptortraffickings waren verstärkt exprimiert. Als ursächlich für die beobachtete verlängerte Hyperalgesie in PAM-defizienten Mäusen konnte die Unterbindung der Internalisierung des TRPV1-Rezeptors nachgewiesen werden. Dieser Effekt ist spezifisch für TRPV1, da der verwandte Ionenkanal TRPA1 durch die PAM-Deletion nicht beeinträchtigt wurde. In der vorliegenden Arbeit konnte so zum ersten Mal gezeigt werden, dass PAM in peripheren nozizeptiven Neuronen über die p38 MAPK-vermittelte Internalisierung von TRPV1 die Dauer der thermischen Hyperalgesie reguliert.
The continuous progress in the structural and functional characterization of aquaporins increasingly attracts attention to study their roles in certain mammalian diseases. Although several structures of aquaporins have already been solved by crystallization, the challenge of producing sufficient amounts of functional proteins still remains. CF (cell free) expression has emerged in recent times as a promising alternative option in order to synthesize large quantities of membrane proteins, and the focus of this report was to evaluate the potential of this technique for the production of eukaryotic aquaporins. We have selected the mouse aquaporin 4 as a representative of mammalian aquaporins. The protein was synthesized in an E. coli extract based cell-free system with two different expression modes, and the efficiencies of two modes were compared. In both, the P-CF (cell-free membrane protein expression as precipitate) mode generating initial aquaporin precipitates as well as in the D-CF (cell-free membrane protein expression in presence of detergent) mode, generating directly detergent solubilized samples, we were able to obtain mg amounts of protein per ml of cell-free reaction. Purified aquaporin samples solubilized in different detergents were reconstituted into liposomes, and analyzed for the water channel activity. The calculated Pf value of proteoliposome samples isolated from the D-CF mode was 133 µm/s at 10°C, which was 5 times higher as that of the control. A reversible inhibitory effect of mercury chloride was observed, which is consistent with previous observations of in vitro reconstituted aquaporin 4. In this study, a fast and convenient protocol was established for functional expression of aquaporins, which could serve as basis for further applications such as water filtration.
Proteins of the Omp85 family are conserved in all kingdoms of life. They mediate protein transport across or protein insertion into membranes and reside in the outer membranes of Gram-negative bacteria, mitochondria, and chloroplasts. Omp85 proteins contain a C-terminal transmembrane β-barrel and a soluble N terminus with a varying number of polypeptide-transport-associated or POTRA domains. Here we investigate Omp85 from the cyanobacterium Anabaena sp. PCC 7120. The crystallographic three-dimensional structure of the N-terminal region shows three POTRA domains, here named P1 to P3 from the N terminus. Molecular dynamics simulations revealed a hinge between P1 and P2 but in contrast show that P2 and P3 are fixed in orientation. The P2-P3 arrangement is identical as seen for the POTRA domains from proteobacterial FhaC, suggesting this orientation is a conserved feature. Furthermore, we define interfaces for protein-protein interaction in P1 and P2. P3 possesses an extended loop unique to cyanobacteria and plantae, which influences pore properties as shown by deletion. It now becomes clear how variations in structure of individual POTRA domains, as well as the different number of POTRA domains with both rigid and flexible connections make the N termini of Omp85 proteins versatile adaptors for a plentitude of functions.

Moderately elevated levels of plasma plant sterols have been suspected to be causally involved in atherosclerosis. The aim of this study was to investigate whether plant sterols and other markers of sterol metabolism predicted all-cause and cardiovascular mortality in participants of the Ludwigshafen Risk and Cardiovascular health (LURIC) study. A total of 1,257 individuals who did not use statins and at baseline had a mean (± SD) age of 62.8 (± 11.0) years were included in the present analysis. Lathosterol, cholestanol, campesterol, and sitosterol were measured to estimate cholesterol synthesis and absorption. The mean (± SD) time of the follow-up for all-cause and cardiovascular mortality was 7.32 (± 2.3) years. All-cause (P = 0.001) and cardiovascular (P = 0.006) mortality were decreased in the highest versus the lowest lathosterol to cholesterol tertile. In contrast, subjects in the third cholestanol to cholesterol tertile had increased all-cause (P < 0.001) and cardiovascular mortality (P = 0.010) compared with individuals in the first tertile. The third campesterol to cholesterol tertile was associated with increased all-cause mortality (P = 0.025). Sitosterol to cholesterol tertiles were not significantly related to all-cause or cardiovascular mortality. The data suggest that high absorption and low synthesis of cholesterol predict increased all-cause and cardiovascular mortality in LURIC participants.
Vibronic (vibrational-electronic) transition is one of the fundamental processes in molecular physics. Indeed, vibronic transition is essential both in radiative and nonradiative photophysical or photochemical properties of molecules such as absorption, emission, Raman scattering, circular dichroism, electron transfer, internal conversion, etc. A detailed understanding of these transitions in varying systems, especially for (large) biomolecules, is thus of particular interest. Describing vibronic transitions in polyatomic systems with hundreds of atoms is, however, a difficult task due to the large number of coupled degrees of freedom. Even within the relatively crude harmonic approximation, such as for Born-Oppenheimer harmonic potential energy surfaces, the brute-force evaluation of Franck-Condon intensity profiles in a time-independent sum-over-states approach is prohibitive for complex systems owing to the vast number of multi-dimensional Franck-Condon integrals. The main goal of this thesis is to describe a variety of molecular vibronic transitions, with special focus on the development of approaches that are applicable to extended molecular systems. We use various representations of Fermi’s golden rule in frequency, time and phase spaces via coherent states to reduce the computational complexity. Although each representation has benefits and shortcomings in its evaluation, they complement each other. Peak assignment of a spectrum can be made directly after calculation in the frequency domain but this sum-over-states route is usually slow. In contrast, computation is considerably faster in the time domain with Fourier transformation but the peak assignment is not directly available. The representation in phase space does not immediately provide physically-meaningful quantities but it can link frequency and time domains. This has been applied to, herein, for example (non-Condon) absorption spectra of benzene and electron transfer of bacteriochlorophyll in the photosynthetic reaction center at finite temperature. This work is a significant step in the treatment of vibronic structure, allowing for the accurate and efficient treatment of complex systems, and provides a new analysis tool for molecular science.
The KADoNiS (Karlsruhe Astrophysical Database of Nucleosynthesis in Stars) project is an online
database (www.kadonis.org) for cross sections relevant to the s-process and the p-process.
The first version was an updated sequel to the previous Bao et al. [1] compilations from 1987
and 2000 for (n; g) cross sections relevant to Big Bang and s-process nucleosynthesis. The first
update, KADoNiS v0.2, was published in 2006 [2]. It contained mainly Maxwellian averaged
(n; g) cross sections relevant to the s-process, and some experimental charged particle induced
reaction relevant to the p-process. After that a second update was presented in 2009 [3].
Recently, we started to collect and review all existing experimental data relevant for p-process
nucleosynthesis and to provide a user-friendly database based on the KADoNiS framework. The
p-process part of the KADoNiS database is currently being extended and will include all available
experimental data from (p; g), (p;n), (p;a), (a,g), (a;n) and (a; p) reactions in or close to the
respective Gamow window.
Die Präkonditionierung mit den bakteriellen Zellwandbestandteilen Lipopolysaccharid (LPS) oder Lipoteichonsäure (LTA) führt in vivo zu einer Reduktion der myokardialen Infarktgröße nach Ischämie und Reperfusion (I/R). Hierbei wird durch die Präkonditionierung u.a. die Akkumulation neutrophiler Granulozyten im Ischämiegebiet während der Reperfusionsphase reduziert und somit einer der wichtigsten Mechanismen bei der Entstehung des Reperfusionsschadens am Herzen vermindert. In dieser Studie bedienten wir uns eines ex vivo Modells nach Langendorff mit regionaler I/R und zellfreier Perfusion. Wir konnten erstmalig eine LTA-Präkonditionierung in einem leukozytenfreien System zeigen und somit demonstrieren, dass die LTA-Präkonditionierung Mechanismen involviert, die unabhängig sind von einer Akkumulation neutrophiler Granulozyten. 24 Stunden nach einer Vorbehandlung der Ratten mit LPS, LTA, Kochsalz und/oder Dexamethason wurden die Herzen entfernt und retrograd mit oxygenierter Krebs-Henseleit-Lösung perfundiert. Die Herzen wurden einer 20-minütigen Ischämie, gefolgt von einer 2-stündigen Reperfusionsphase, unterzogen. Das Infarktrisikogebiet (Evans-Blue-Färbung) und das Infarktgebiet (pNBT-Färbung) wurden planimetrisch bestimmt. Die ischämische Präkonditionierung (IPC) wurde als Positivkontrolle unseres Modells verwendet. LTA- und LPS-Präkonditionierung führten - ebenso wie IPC - bei gleicher Dosierung in vergleichbarem Umfang zu einer signifikanten Reduktion der Infarktgröße. Dieser Effekt konnte durch Vorbehandlung mit Dexamethason vollständig aufgehoben werden, so dass gefolgert werden kann, dass die Protektion durch LPS bzw. LTA. von der Modulation der inflammatorischen Vorgänge im Endothel und Myokard mit abhängt.
Surface measurements of aerosol and ice nuclei (IN) at a Central European mountain site during an episode of dust transport from the Sahara are presented. Ice nuclei were sampled by electrostatic precipitation on silicon wafers and were analyzed in an isothermal static vapor diffusion chamber. The transport of mineral dust is simulated by the Eulerian regional dust model DREAM. Ice nuclei and mineral dust are significantly correlated, in particular IN number concentration and aerosol surface area. The ice nucleating characteristics of the aerosol as analyzed with respect to temperature and supersaturation are similar during the dust episode than during the course of the year. This suggests that dust may be a main constituent of ice nucleating aerosols in Central Europe.

Durch die steigende Bedeutung von multimedialen Lernmaterialien in der Lehre und in der Wirtschaft, wachsen die Anforderungen, die an die Herstellung der Materialien gestellt werden. Verwaltung, Archivierung und Wiederverwendbarkeit sind die zentralen Begriffe um die Effektivität von multimedialen Lernmaterialien zu steigern und damit auch kommerziellen Erfolg zu erzielen. Es ist effektiv wenn Lernmaterialien ein Thema nicht erneut aufarbeiten, wenn es bereits ausreichend Materialien dazu gibt. Vorhandene Materialien können genutzt werden und sparen somit Ressourcen, die ansonsten zur Herstellung neuer Materialien genutzt werden müssten. In dieser Arbeit werden Metadaten als Mittel eingesetzt, um die Effektivität zu steigern. Dabei soll vor allem der Mehrwert durch die Verwendung von Metadaten deutlich gemacht werden. Eine Analyse aktueller Autorensysteme soll zeigen, wie Metadaten bereits eingesetzt werden und wo Probleme existieren. Die Stärken und Schwächen der untersuchten Autorensysteme werden für die Anforderungsanalyse einer Erweiterung des Autorensystems LernBar verwendet. Um den Mehrwert der Metadaten deutlich zu machen, soll der gesamte Autorenprozess innerhalb der LernBar erweitert werden. Das heißt, dass der gesamte Ablauf, vom Erfassen bis zum Verwenden der Metadaten, abgebildet wird. Im Fokus der Erweiterung steht die Unterstützung des Autors bei der Erstellung von Lernmaterialien. Vorlagen und automatisierte Vorgänge dienen der Bedienbarkeit der neuen Funktionen.
Background: Many disabling human retinal disorders involve the central retina, particularly the macula. However, the commonly used rodent models in research, mouse and rat, do not possess a macula. The purpose of this study was to identify small laboratory rodents with a significant central region as potential new models for macular research.
Methodology/Principal Findings: Gerbillus perpallidus, Meriones unguiculatus and Phodopus campbelli, laboratory rodents less commonly used in retinal research, were subjected to confocal scanning laser ophthalmoscopy (cSLO), fluorescein and indocyanine green angiography, and spectral-domain optical coherence tomography (SD-OCT) using standard equipment (Heidelberg Engineering HRA1 and Spectralis™) adapted to small rodent eyes. The existence of a visual streak-like pattern was assessed on the basis of vascular topography, retinal thickness, and the topography of retinal ganglion cells and cone photoreceptors. All three species examined showed evidence of a significant horizontal streak-like specialization. cSLO angiography and retinal wholemounts revealed that superficial retinal blood vessels typically ramify and narrow into a sparse capillary net at the border of the respective area located dorsal to the optic nerve. Similar to the macular region, there was an absence of larger blood vessels in the streak region. Furthermore, the thickness of the photoreceptor layer and the population density of neurons in the ganglion cell layer were markedly increased in the visual streak region.
Conclusions/Significance: The retinal specializations of Gerbillus perpallidus, Meriones unguiculatus and Phodopus campbelli resemble features of the primate macula. Hence, the rodents reported here may serve to study aspects of macular development and diseases like age-related macular degeneration and diabetic macular edema, and the preclinical assessment of therapeutic strategies.
The display of foreign polypeptides and proteins on the surface of viruses or cells provides an important tool for the engineering of biomolecules and the analysis of their interactions with binding partners. The most extensively used display platform is the coat protein of the filamentous bacteriophage (Smith, 1985). Phage display libraries have often been selected for polypeptides, e.g. single chain (sc) antibodies that bind to a protein of interest, but in vivo selection could only be demonstrated for peptides so far. An alternative display platform is the retrovirus murine leukemia virus (MLV). Here, polypeptides are displayed at the N-terminus of the viral envelope glycoprotein. Proof of principle for this platform was demonstrated for protease substrate libraries, which can be selected through coupling proteolytic activation with viral infectivity (Buchholz et al., 1998). Selection of the library CX4A on living cells resulted in viruses with more than three orders of magnitude improved spreading efficiency through tumor cells (Hartl et al., 2005). Also scAb libraries have recently been displayed and selected using retroviruses (Urban et al., 2005). The library scFvlibxMo displays the repertoire of phage display preselected sc antibodies for laminin-1 binding. The retrovirus based selection process resulted in laminin-specific sc antibodies with improved expression levels in mammalian cells.
This thesis describes the in vivo (i.e. in mouse tumor models) selection of the C-X4-A and scFvlibxMo for tumor homing upon systemic delivery.
For selection of the protease substrate library C-X4-A a subcutaneous tumor was induced in SCID mice followed by three systemic injections of the library. The selection process was monitored over a period of 34 days. After the incubation period mice were sacrificed and virus load in organs and tumor determined. PCR analysis after 34 days showed that virus from the library had preferentially infected the tumor. Sequence analysis showed the selection of protease substrates with the most prominent one with a frequency of over 65%. The four most prominent protease substrate variants where reconstituted into the original viral backbone for further investigation (C-SK-A, C-HI-A, C-HM-A and C-HS-A). Interestingly, these viruses exhibited a reduced spreading capacity in vitro on HT1080 cells as compared to the C-AK-A virus, which had previously been selected on HT1080 cells. When assayed for tumor homing, however, viruses C-HI-A and C-HS-A had clearly improved in comparison to C-AK-A. Tumor tissue had been infected at rates of over 55% while virus load of extratumoral organs was very low (infection rates <0.7 for C-HS-A and <0.02 for C-HI-A). Tumor targeting capacity had thus been improved over 10-fold by the in vivo selection of the C-X4-A library.
The experimental set up for the in vivo selection of the scFvlibxMo library was performed according to that of the C-X4-A library. Fingerprint analysis of the selected viruses that infected tumor tissue resulted in the identification of seven antibody variants showing unique CDR3 sequences. Two prominent clones (M49T-A and M49T-B) were cloned back into the MoMLV genome for further analysis of the reconstituted viruses. While variant B bound laminin-1 efficiently, variant A was unable to do so, although it was selected at highest frequency (76%). Both reconstituted viruses were equally well infectious and spread through HT1080rec1 cells at a similar efficiency as MoMLV. In an in vivo competition experiment the selected viruses clearly out-competed a laminin-1 binding reference virus L36xMo for tumor homing. To understand the molecular driving forces behind the in vivo selection process the epitope of the selected scFv M49T-A was identified using a phage peptide library approach. In silico analysis led to the identification of a small group of possible antigens, including tenascin, fibronectin and collagen.
The data described in this thesis demonstrate that the retrovirus display platform is capable of allowing the in vivo selection of protease substrates and scFvs. Notably, the replication competence of the system introduced an additional level of complexity to the library. The performed in vivo selections significantly enhanced tumor tropism. Selective infection of tumor cells combined with transfer of anti-tumoral genes is an attractive strategy for cancer therapy being in focus of current research. The viruses selected in this thesis build prime candidates for targeted retrovirus based tumor therapy.
Aufbau der Arbeit Auf der Grundlage einer Definition von Integrationsprozessen bzw. Integrationszuständen (Kap. 2.1) werden Heterogenitätsmerkmale diskutiert, die für Anerkennungsunterschiede zwischen Ingroup und Outgroup verantwortlich sein können (Kap. 2.1.2). Anschließend werden Integrations- und Desintegrationsmechanismen moderner Gesellschaften aufgezeigt und Erkenntnisse der Vorurteilsforschung zur Verbesserung der Outgroup-Anerkennung erörtert (Kap. 2.1.3 und 2.1.4). Im darauffolgenden Kapitel 2.2 werden Formen des Sports im Hinblick auf deren Integrations- und Desintegrationspotenziale diskutiert. Kapitel 2.3 beschäftigt sich zunächst mit der grundlegenden pädagogischen Rahmung des erziehenden Sportunterrichts, stellt einzelne Integrationsansätze vor und legt dabei den Fokus auf die Frage der bewussten oder unbewussten Auseinandersetzung mit Heterogenität. Da Kooperatives Lernen als eine erfolgversprechende Methode für gelingende Integration gehandelt wird und diese Methode zudem Integrationsmechanismen sowie Kontakthypothese berücksichtigt, wird sie in Kapitel 2.4 ausführlich beschrieben. Nachdem die wichtigsten Theorien zu heterogenitätsbedingten Anerkennungsunterschieden sowie integrationspädagogischen Überlegungen im Sportunterricht behandelt worden sind, schließt der Theorieteil mit der Ableitung der Fragestellungen (Kap. 2.5). Fragestellungen: In der soziologischen Untersuchung wird der Frage nach den tatsächlich desintegrierenden Heterogenitätsmerkmalen im Sportunterricht nachgegangen. In der pädagogischen Untersuchung wird erforscht, ob Kooperatives Lernen im Sportunterricht in der Lage ist, den selektiven Einfluss der desintegrierenden Heterogenitätsmerkmale abzubauen.
Wassergefiltertes Infrarot A (wIRA) ist eine spezielle Form der Wärmestrahlung mit hohem Penetrationsvermögen ins Gewebe bei geringer thermischer Oberflächenbelastung. wIRA entspricht dem Großteil der Sonnenwärmestrahlung, die in gemäßigten Klimazonen die Erdoberfläche wasserdampfgefiltert erreicht. wIRA steigert die drei energetisch für die Wundheilung wichtigen Faktoren Temperatur, Sauerstoffpartialdruck und Durchblutung im Gewebe. wIRA mindert Schmerzen, Entzündung und Wundsekretion. Entsprechend kann wIRA sehr gut zur Verbesserung der Wundheilung bei akuten und chronischen Wunden eingesetzt werden.
Poster presentation at 5th German Conference on Cheminformatics: 23. CIC-Workshop Goslar, Germany. 8-10 November 2009 We demonstrate the theoretical and practical application of modern kernel-based machine learning methods to ligand-based virtual screening by successful prospective screening for novel agonists of the peroxisome proliferator-activated receptor gamma (PPARgamma) [1]. PPARgamma is a nuclear receptor involved in lipid and glucose metabolism, and related to type-2 diabetes and dyslipidemia. Applied methods included a graph kernel designed for molecular similarity analysis [2], kernel principle component analysis [3], multiple kernel learning [4], and, Gaussian process regression [5]. In the machine learning approach to ligand-based virtual screening, one uses the similarity principle [6] to identify potentially active compounds based on their similarity to known reference ligands. Kernel-based machine learning [7] uses the "kernel trick", a systematic approach to the derivation of non-linear versions of linear algorithms like separating hyperplanes and regression. Prerequisites for kernel learning are similarity measures with the mathematical property of positive semidefiniteness (kernels). The iterative similarity optimal assignment graph kernel (ISOAK) [2] is defined directly on the annotated structure graph, and was designed specifically for the comparison of small molecules. In our virtual screening study, its use improved results, e.g., in principle component analysis-based visualization and Gaussian process regression. Following a thorough retrospective validation using a data set of 176 published PPARgamma agonists [8], we screened a vendor library for novel agonists. Subsequent testing of 15 compounds in a cell-based transactivation assay [9] yielded four active compounds. The most interesting hit, a natural product derivative with cyclobutane scaffold, is a full selective PPARgamma agonist (EC50 = 10 ± 0.2 microM, inactive on PPARalpha and PPARbeta/delta at 10 microM). We demonstrate how the interplay of several modern kernel-based machine learning approaches can successfully improve ligand-based virtual screening results.
With the help of miniaturized GPS recorders I recorded 167 tracks of 48 individual pigeons during their flight from 6 different sites around Frankfurt. The experiments consisted of two main series of repeated releases from two sites 30 km north and south from the pigeons' home loft. From the site in the south the pigeons homed 12 times and from the site in the north 16 times. After the final release from these sites, the pigeons were released at 60 km distance from home. These additional sites were selected so that the pigeons would presumably fly over the previous release site with which they were highly familiar. After conclusion of the main series two additional releases were performed, one within the magnetic anomaly of the Vogelsberg and one in a magnetically quiet region. To make these releases comparable, both release sites were selected so that the distance from the home loft was 40 km. All data obtained during these experiments were subjected to a threefold analysis, mostly based on methods that I had developed by myself or adapted for this specific study. In the first step, data were analyzed traditionally, evaluating variables similar to those that can be found in current literature. I therefore calculated values that correspond to those obtained by visual observation, like virtual vanishing bearings and intervals after one minute and after 2.5 km. Additionally I calculated the efficiency of the flights and efficiencies for specific portions of each flight, to derive variables that describe the behavior after vanishing. In the second step, which served also as a preparation for the mathematical analysis, the flight of the pigeons was separated into distinctive phases of the flight by the so-called points of decision. The flight of the pigeon can usually be separated into an initial phase of flying about, a departure and/or final homing phase. In more complex cases, however, several points of decision and a multitude of intermediary phases can be defined. Yet, the initial phase, the departure phase and the final homing phase can be defined for all tracks and therefore have been selected as appropriate candidates for a thorough analysis. In the last step I employed the so-called method of time lag embedding to reconstruct the underlying navigational process of the pigeons' homing flight. This method is based on the principles of chaos theory and is regularly employed for the analysis of dynamic systems. Its application allows the reconstruction of the underlying processes from experimentally recorded data without any a priori knowledge of the underlying system itself. For these reconstructed systems I calculated characteristic properties which are unique for each system. These are the so-called correlation dimension, describing the complexity of the system, and the so-called largest Lyapunov exponent, describing its predictability. Based on the knowledge gathered from these reconstructions, I used a variation of the previous methods to identify navigational phases, by calculating the correlation dimension as a sliding mean over the complete track. From these data I then derived further characteristics of the underlying process, such as its precision and differences in complexity depending on the pigeon's current position. ...
Mitte des 19. Jahrhunderts demonstrierte John Snow anhand differenzierter Beobachtungen zur Cholera in London, wie epidemiologisches Wissen und gezielte Maßnahmen zur Bewältigung öffentlicher Gesundheitsprobleme beitragen können. Rund 150 Jahre später sieht sich die Bevölkerung einem stetig wachsenden globalen Güter- und Personenverkehr gegenüber, welcher auch Krankheitserregern eine interkontinentale Ausbreitung innerhalb weniger Stunden ermöglicht, wie eindrucksvoll am Beispiel SARS im Jahre 2003 deutlich wurde. Nationale Beispiele, allen voran die Salmonellen-Epidemie in Fulda im Jahre 2007, zeigen, welche bedeutungsvolle Rolle die Infektionsepidemiologie und die -hygiene auch im 21. Jahrhundert einnimmt. Das frühzeitige Erkennen und ein effizientes Eingreifen durch die Öffentlichen Gesundheitsbehörden sind zur Eindämmung einer Epidemie unabdingbar. Die Verknüpfung medizinischer und geographischer Daten kann Beides wesentlich beschleunigen und ermöglicht die frühzeitige Erkennung eskalierender Infektionsherde. Ziel der vorliegenden Pilotstudie ist die Entwicklung einer Schnittstelle zur Implementierung und Analyse meldepflichtiger Infektionskrankheiten in einem geomedizinischen Informationssystem. Erstmals im Öffentlichen Gesundheitsdienst wird diese Verknüpfung technisch mittels eines Geoinformationssystems realisiert, welches die Georeferenzierung mithilfe von Regionalidentifikationsnummern und der anschließende Visualisierung der im Gesundheitsamt anfallenden krankheitsbezogenen Daten ermöglicht. Der Datentransfer von dem im Amt für Gesundheit genutzten Datenbankprogramm Gumax® zu dem im Vermessungsamt der Stadt Frankfurt am Main probaten Geoinformationssystem Office-GIS gelingt über einen SQL-Server, einem Datenbankmanagementsystem, welches das Speichern, Bearbeiten und Analysieren vergleichsweise großer Datenmengen ermöglicht. Anschließend können Meldeort und Wohnort des an einer nach §§ 6, 7 IfSG meldepflichtigen Infektionserkrankung Erkrankten in der Stadtplan-, Liegenschaftskarte oder Luftbildaufnahme visualisiert werden. Hierüber lassen sich zudem personen- und objektbezogene Krankheitsquellen (z. B. Restaurant, Schule, Kindergarten, Krankenhaus) eruieren. Diese Daten können effizient genutzt werden, um schnell und dezidiert in ein Krankheitsgeschehen eindämmend eingreifen zu können. Mit diesem System könnten auch bioterroristische Anschläge wesentlich schneller erkannt werden, da die Ausbreitungsmodalitäten beispielsweise vom verwendeten Agens, meteorologischen, tageszeitlichen und demographischen Gegebenheiten abhängen. Diesen zusätzlichen Größen soll in erweiterten technischen Realisationen dieses Systems Rechnung getragen werden.
Influenza A (H1N1) 2009 : impact on Frankfurt in due consideration of health care and public health

Background: In April 2009 a novel influenza A H1N1/2009 virus was identified in Mexico and in the United States which quickly spread around the world. Most of the countries established infection surveillance systems in order to track the number of (laboratory-confirmed) H1N1 cases, hospitalizations and deaths. Methods: The impact of the emergence of the novel pandemic (H1N1) 2009 virus on Frankfurt was statistically evaluated by the Health Protection Authority, City of Frankfurt am Main. Vaccination rates of the health care workers (HCWs) of the University Hospital Frankfurt were measured by the Occupational Health Service. Results: Although the virulence of pandemic (H1N1) 2009 seems to be comparable with seasonal influenza, a major patient load and wave of hospital admissions occurred in the summer of 2009. Even though the 2009 vaccination rate of the University Hospital Frankfurt (seasonal influenza [40.5%], swine flu [36.3%]) is better than the average annual uptake of influenza vaccine in the German health care system (approximately 22% for seasonal and 15% for swine flu), vaccination levels remain insufficient. However, physicians were significantly (p < 0.001) more likely to have been vaccinated against swine flu and seasonal influenza than nurses. Conclusions: The outbreak of the pandemic (H1N1) 2009 in April 2009 provided a major challenge to health services around the world. Nosocomial transmission of H1N1/2009 has been documented. Present experience should be used to improve pandemic preparedness plans and vaccination programs ought to target as many HCWs as possible.
Determinanten der Langzeitprognose von Patienten mit implantierbaren Cardioverter-Defibrillatoren

Die Effektivität der ICD-Therapie in der Verhinderung des Plötzlichen Herztodes ist unbestritten. Implantierbare Defibrillatoren können ventrikuläre Tachykardien und Kammerflimmern mit hoher Sicherheit beenden und damit den Plötzlichen Herztod mit sehr hoher Wahrscheinlichkeit verhindern. Die vorliegende Studie untersuchte Determinanten der Langzeitprognose von Patienten nach ICD-Implantation. Dabei zeigte sich in unserer Studie bei einer jährlichen Mortalität von 7,2% eine Sterblichkeit am Plötzlichen Herztod von < 1% pro Jahr, was den Nutzen des ICD unterstreicht. Bei der Analyse der Mortalitätsparameter waren ein hohes Alter der Patienten, fortgeschrittene Herzinsuffizienz mit entsprechendem NYHA-Stadium sowie eine deutlich eingeschränkte linksventrikuläre Ejektionsfraktion gute Vorhersagevariablen für die Prognose nach ICD-Implantation mit hohem prädiktiven Wert für die Mortalität. Ebenso scheint die ICD-Indikation ein wichtiger Parameter für die Langzeitprognose zu sein (sekundärprophylaktisch ICD-implantierte Patienten verstarben signifikant häufiger als Patienten, die zur Primärprophylaxe mit einem ICD versorgt wurden), primärprophylaktische Patienten stellen ein Patientenkollektiv mit per se besserer Prognose dar. Bezüglich der Auswertung des Auftretens adäquater Therapien erwiesen sich männliches Geschlecht, hohes Alter, Einnahme antiarrhythmischer Medikation sowie die sekundärprophylaktische ICD-Indikation als bedeutsame Parameter für eine erhöhte ICD-Interventions-Wahrscheinlichkeit. Zur Frage der prognostischen Bedeutung adäquater Therapien zeigte sich, dass im Auftreten adäquater Therapien ebenfalls eine gute Vorhersagevariable für die Langzeitprognose zu sehen ist. Die Auftretenswahrscheinlichkeit von adäquaten ICD-Therapien korrelierte jedoch nichtsignifikant mit dem Plötzlichen Herztod und somit scheinen vorhergehende ICD-Interventionen kein erhöhtes Risiko für diesen darzustellen. Die technische Weiterentwicklung hat in den letzten Jahren zu einer Größenreduktion der ICD´s, zur Vereinfachung der Systemimplantation mit verschwindend geringen peri- und postoperativen Komplikations- und Mortalitätsraten sowie durch die Verlängerung der Systemlebensdauer zum zunehmenden Einsatz von implantierbaren Defibrillatoren als bevorzugte Therapieform mit unbestrittener Effektivität in der Terminierung lebensbedrohlicher Arrhythmien, geführt. Trotzdem wären zukünftig prospektive Studien zur weiteren Risikostratifizierung und Evaluation möglicher diagnostischer Marker zur noch genaueren Abgrenzung von Hochrisikopatienten wünschenswert. Zukünftiges Ziel sollte es zudem sein, diejenigen Patientenpopulationen sicher zu identifizieren, die zwar ein geringeres individuelles Risiko haben dem Plötzlichen Herztod zu erliegen, jedoch absolut gesehen häufiger daran versterben.

X-Ray quality crystals of H2C=C(CH2SitBu2-NH-SiPhtBu2)2 (monoclinic, space group C2/c, Z = 4) were grown from a benzene solution at ambient temperature, whereas single crystals of H2C=C(CH2SitBu2-NH-SitBu3)2 (monoclinic, space group P21, Z = 2) were obtained by recrystallization from THF.

Dieser Sammelband widmet sich sehr verschiedenen Aspekten der Konzilien von Pisa, Konstanz und Basel und des Konziliarismus des 15. Jahrhunderts. In seiner Zusammenfassung am Ende muss Werner Maleczek gestehen, dass eine thematische Gliederung der Aufsätze kaum möglich ist. Der "vielgestaltige Reichtum" der Beiträge lässt diese Gliederung nicht zu. In ihrer thematischen Unterschiedlichkeit ist ihnen aber doch eine gemeinsame Spannung eigen. Es ist schon erstaunlich, einen Sammelband zu Themen zu erhalten, über die eine nahezu unüberschaubare Fülle an Literatur erschienen ist, und bei der Lektüre gewissermaßen den Eindruck zu gewinnen, dass es sich um ein neues Forschungsfeld handelt. Es wird immer wieder nach Grundsätzlichem gefragt. Fast könnte man sagen, dass die Fragen "Was ist ein Konzil?" und "Was ist Konziliarismus?" die Hauptthemen des Buches sind. Auf diese Weise enthält der Band, aus meiner Sicht, sehr viele Gedanken, die das Potential haben, die Forschung zu alten Themen zu erneuern. Auf diesen Aspekt möchte ich mich konzentrieren, auch, wenn das heißt, dass Einiges oder sogar Vieles Erwähnenswertes übergangen werden muss. ...
Entzündliche Herzerkrankungen betreffen kombiniert oder isoliert den Herzmuskel und dessen Hülle. Endo-, Myo- und/oder Perikarditiden haben viele verschiedene Ursachen. Sie verlaufen als akute oder chronische Erkrankung. Neben Viren, die gegenwärtig als auslösende Agentien dominieren, sind weiterhin Bakterien, Pilze und Parasiten anzuführen. Autoimmunologische Prozesse sowie bestimmte Therapeutika, z.B. Cocain, gelten als Auslöser nicht infektiöser Myokarditiden. In 25% der Fälle findet sich bei bestehender Myokarditis eine Perikardbeteiligung. Nachfolgend sollen wichtige mikrobiologische Erreger und deren Nachweismöglichkeiten vorgestellt werden, die im Zusammenhang mit einer Myo- und/oder Perikarditis stehen.
Qualitative und quantitative serologische Verfahren können durch Interferenzen gestört sein. Wir konnten in einem exemplarischen Fall anhand des Influenza A/H1N1v-Hämagglutinationshemmtests (H1N1-HHT) zeigen, dass auch Hyposensibilisierungstherapie und Vakzination zu Interaktionen in der serologischen Diagnostik führen und die Aussagekraft des H1N1-HHT massiv beeinträchtigen. Vor dem Hintergrund, dass Hyposensibilisierung und Vakzination im Klinik- und Praxisalltag häufig erbrachte Leistungen darstellen, erscheint dieser Umstand berichtenswert.
In Europa zählen Viren zu den häufigsten Verursachern einer Myokarditis. Im Unterschied zur Perikarditis ist die Symptomatik der Myokarditis oft uncharakteristisch und erfordert den Einsatz von Laboruntersuchungen. Die Abklärung der Virusätiologie begnügt sich meist mit dem Nachweis einer zeitgleich ablaufenden Infektionskrankheit (Plausibilitätsdiagnose). Zur direkten Virusdetektion ist die Entnahme einer Herzbiopsie erforderlich. An diesem Material kann das Virus mittels immunhistologischer und molekularbiologischer Methoden unter gleichzeitiger Beurteilung des inflammatorischen Prozesses nachgewiesen werden. Der Einsatz der verschiedenen Untersuchungsmethoden richtet sich nach dem Kosten-Nutzen-Verhältnis.
Eifrig, sorgfältig und beharrlich fügt Jacques Paviot mit seinen Publikationen seit Jahren Stein auf Stein, um so eines Tages das große Gebäude einer Darstellung französischer Kreuzzugspläne und -politik im späten Mittelalter vollenden zu können; wichtige Teile hierzu hat er bereits mit seinen Studien und Editionen zur croisade bourguignonne und da insbesondere zu den spektakulären Projekten und Unternehmen aus der Zeit Philipps d. In der recht strikt reglementierten Wissenschafts- und Universitätslandschaft Frankreichs gibt es ein Refugium akademischer Freiheit und Exzellenz, eine Art Super-"Institute for Advanced Study" und dies mit einer bis zu Franz I. und Guillaume Budé in das Jahr 1530 reichenden Tradition: das bereits früh an heutiger Stätte im Pariser Quartier latin ansässige Collège de France. Dessen gegenwärtig etwas über 50 Professoren – am Anfang standen lediglich "lecteurs royaux" für die klassischen Sprachen – obliegt eine einzige Aufgabe, ob es sich nun um Mathematiker, Naturwissenschaftler, Informatiker, Philosophen, Soziologen, Historiker und Philologen, um Astrophysiker, Assyrologen oder Psychologen handelt: das Wissen, wie es entsteht, zu lehren ("enseigner le savoir en train de se faire"). Solche Lehre erfolgt gratis et publice, sie führt weder zu Prüfungen noch zu Abschlüssen; im Idealfall eint Lehrende und Lernende reiner amor scientiae. Entscheidend für eine Berufung sind Gewicht und Originalität der wissenschaftlichen Persönlichkeit; deshalb können bei Emeritierung, Weggang oder Tod freigewordene Lehrstühle auf Initiative der kooptierenden Professorenschaft dem Arbeitsgebiet der gewünschten Kandidaten entsprechend umgewidmet werden. ...
If insufficiently treated, Lyme borreliosis can evolve into an inflammatory disorder affecting skin, joints, and the CNS. Early innate immunity may determine host responses targeting infection. Thus, we sought to characterize the immediate cytokine storm associated with exposure of PBMC to moderate levels of live Borrelia burgdorferi. Since Th17 cytokines are connected to host defense against extracellular bacteria, we focused on interleukin (IL)-17 and IL-22. Here, we report that, despite induction of inflammatory cytokines including IL-23, IL-17 remained barely detectable in response to B. burgdorferi. In contrast, T cell-dependent expression of IL-22 became evident within 10 h of exposure to the spirochetes. This dichotomy was unrelated to interferon-gamma but to a large part dependent on caspase-1 and IL-1 bioactivity derived from monocytes. In fact, IL-1beta as a single stimulus induced IL-22 but not IL-17. Neutrophils display antibacterial activity against B. burgdorferi, particularly when opsonized by antibodies. Since neutrophilic inflammation, indicative of IL-17 bioactivity, is scarcely observed in Erythema migrans, a manifestation of skin inflammation after infection, protective and antibacterial properties of IL-22 may close this gap and serve essential functions in the initial phase of spirochete infection.
Background In October 2007, the working group CEN/TC 216 of the European Committee for standardisation suggested that the Sabin oral poliovirus vaccine type 1 strain (LSc-2ab) presently used for virucidal tests should be replaced by another attenuated vaccine poliovirus type 1 strain, CHAT. Both strains were historically used as oral vaccines, but the Sabin type 1 strain was acknowledged to be more attenuated. In Germany, vaccination against poliomyelitis was introduced in 1962 using the oral polio vaccine (OPV) containing Sabin strain LSc-2ab. The vaccination schedule was changed from OPV to an inactivated polio vaccine (IPV) containing wild polio virus type 1 strain Mahoney in 1998. In the present study, we assessed potential differences in neutralising antibody titres to Sabin and CHAT in persons with a history of either OPV, IPV, or OPV with IPV booster. Methods Neutralisation poliovirus antibodies against CHAT and Sabin 1 were measured in sera of 41 adults vaccinated with OPV. Additionally, sera from 28 children less than 10 years of age and immunised with IPV only were analysed. The neutralisation assay against poliovirus was performed according to WHO guidelines. Results The neutralisation activity against CHAT in adults with a complete OPV vaccination series was significantly lower than against Sabin poliovirus type 1 strains (Wilcoxon signed-rank test P < 0.025). In eight sera, the antibody titres measured against CHAT were less than 8, although the titre against Sabin 1 varied between 8 and 64. Following IPV booster, anti-CHAT antibodies increased rapidly in sera of CHAT-negative adults with OPV history. Sera from children with IPV history neutralised CHAT and Sabin 1 strains equally. Conclusion The lack of neutralising antibodies against the CHAT strain in persons vaccinated with OPV might be associated with an increased risk of reinfection with the CHAT polio virus type 1, and this implies a putative risk of transmission of the virus to polio-free communities. We strongly suggest that laboratory workers who were immunised with OPV receive a booster vaccination with IPV before handling CHAT in the laboratory.
Es werden 66 Neu- oder Wiederfunde sowie nicht publizierte Funde von Flechten und flechtenbewohnenden Pilzen in Hessen gemeldet und kurz kommentiert. Sechs Arten sind zugleich Neufunde für Deutschland: Absconditella pauxilla, Acremonium antarcticum, Calicium adaequatum, Psammina inflata, Ramonia chrysophaea und Tubeufia heterodermiae. Die hessische Artenliste umfasst mittlerweile annähernd 1200 Flechten und flechtenbewohnende oder flechtenähnliche Pilze.
