1987 überraschte der chinesische Künstler Xu Bing sein Pekinger Publikum mit einem Set von vier Büchern, die im Stil der kanonischen Bücher gestaltet waren und über tausendzweihundert chinesische Schriftzeichen enthielten. Obwohl auf den ersten Blick den alten chinesischen Zeichen täuschend ähnlich, unterschieden sie sich jedoch von ihnen in einigen wenigen Details, die entscheidend für das Verständnis waren. Xu Bings Idee bestand darin, einem alten Wörterbuch reale Zeichen zu entnehmen, sie in ihre Grundbestandteile zu zerlegen und sie wieder zusammensetzen, jedoch nicht so, wie sie vorher waren, sodass sie ihrer Bedeutung beraubt wurden. Angesichts dieser gekonnt inszenierten und verwirrenden Lektüreerfahrung stellt sich die Frage: Ist das überhaupt Literatur, wo es doch nichts zu lesen gibt? Diese Frage mag ungewöhnlich klingen und nicht wenige werden sie vielleicht sogar für falsch gestellt halten. Schließlich gilt Xu Bing als Künstler und nicht als Schriftsteller und seine ausgestellten Bücher sind in dem Ausstellungsraum eines Museums zu finden und nicht in einer Bibliothek. Und dennoch, so die Hypothese dieser Überlegungen, findet man in seinen Büchern etwas zur Darstellung gebracht, was nach Foucault so etwas ist wie der unsagbare Kern der Literatur. In Xu Bings Ausstellung geht es um die sichtbare Manifestation dessen, was Literatur im sprachontologischen Sinne begründet, um ihre Existenzbedingung: das Nichtsignifikative im Zeichen, das die Signifikation erst ermöglicht. In Analogie zu Blanchots Begriff "desoeuvrement" (vgl. Blanchot 1955, 48 f) könnte man von einer 'désignification' sprechen: Durch winzige Veränderungen am Gefüge der Striche wird dem Zeichen der Sinn entzogen. Doch wie lässt sich der Bezug dieses unlesbaren Buches, das aufgrund seiner Unlesbarkeit nur noch als Ausstellungsobjekt taugt, zum literarischen Buch herstellen?
Motion analysis in the field of dentistry : a kinematic comparison of dentists and orthodontists

Objectives: To conduct a kinematic comparison of occupational posture in orthodontists and dentists in their workplace.
Design: Observational study.
Setting: Dentist surgeries and departments of orthodontics at university medical centres in Germany.
Participants: A representative sample of 21 (10 female, 11 male) dentists (group G1) and 21 (13 female, 8 male) orthodontists (G2) with one male dropout in G2.
Outcome measures: The CUELA (computer-assisted acquisition and long-term analysis of musculoskeletal loads) system was used to analyse occupational posture. Parallel to the recording through the CUELA system, a software-supported analysis of the activities performed (I: treatment; II: office; III: other activities) was carried out. In line with ergonomic standards the measured body angles are categorised into neutral, moderate and awkward postures. Activities between the aforementioned groups are compared using the stratified van Elteren U test and the Wilcoxon–Mann–Whitney U test. All p values are subject to the Bonferroni–Holm correction. The level of significance is set at 5%.
Results: The percentage of time spent on activities in categories I–II–III was as follows: dentists 41%–23%–36% and orthodontists 28%–37%–35%. The posture analysis of both groups showed, for all percentiles (P5–95), angle values primarily in the neutral or moderate range. However, depending on the activity performed, between 5% and 25% of working hours were spent in unfavourable postures, especially in the head-and-neck area. Orthodontists have a greater tendency than dentists to perform treatment activities with the head and torso in unfavourable positions. The statistically significant differences between the two groups with regard to the duration and the relevance of the activities performed confirm this assumption for all three categories (p<0.01, p<0.05).
Conclusions: Generally, both groups perform treatment activities in postures that are in the neutral or medium range; however, dentists had slightly more unfavourable postures during treatment for a greater share of their work day.
The acoustic startle response (ASR) and its modulation by non-startling prepulses, presented shortly before the startle-eliciting stimulus, is a broadly applied test paradigm to determine changes in neural processing related to auditory or psychiatric disorders. Modulation by a gap in background noise as a prepulse is especially used for tinnitus assessment. However, the timing and frequency-related aspects of prepulses are not fully understood. The present study aims to investigate temporal and spectral characteristics of acoustic stimuli that modulate the ASR in rats and gerbils. For noise-burst prepulses, inhibition was frequency-independent in gerbils in the test range between 4 and 18 kHz. Prepulse inhibition (PPI) by noise-bursts in rats was constant in a comparable range (8–22 kHz), but lower outside this range. Purely temporal aspects of prepulse–startle-interactions were investigated for gap-prepulses focusing mainly on gap duration. While very short gaps had no (rats) or slightly facilitatory (gerbils) influence on the ASR, longer gaps always had a strong inhibitory effect. Inhibition increased with durations up to 75 ms and remained at a high level of inhibition for durations up to 1000 ms for both, rats and gerbils. Determining spectral influences on gap-prepulse inhibition (gap-PPI) revealed that gerbils were unaffected in the limited frequency range tested (4–18 kHz). The more detailed analysis in rats revealed a variety of frequency-dependent effects. Gaps in pure-tone background elicited constant and high inhibition (around 75%) over a broad frequency range (4–32 kHz). For gaps in noise-bands, on the other hand, a clear frequency-dependency was found: inhibition was around 50% at lower frequencies (6–14 kHz) and around 70% at high frequencies (16–20 kHz). This pattern of frequency-dependency in rats was specifically resulting from the inhibitory effect by the gaps, as revealed by detailed analysis of the underlying startle amplitudes. An interaction of temporal and spectral influences, finally, resulted in higher inhibition for 500 ms gaps than for 75 ms gaps at all frequencies tested. Improved prepulse paradigms based on these results are well suited to quantify the consequences of central processing disorders.
The chloroplast phosphorylation network is important for posttranslational regulation of photosynthetic complexes, gene expression and metabolic pathways. In mass-spectrometric analyses a lot of putative phosphorylation targets have been found but these data need to be confirmed and brought into a physiological context. Here, we present a current protocol to quantify the phosphorylation state of thylakoid proteins and an in situ method to verify putative substrates for thylakoid associated kinases.
One hallmark of MLL-r leukemia is the highly specific gene expression signature indicative for commonly deregulated target genes. An usual read-out for this transcriptional deregulation is the HOXA gene cluster, where upregulated HOXA genes are detected in MLL-r AML and ALL patients. In case of t(4;11) leukemia, this simple picture becomes challenged, because these patients separate into HOXAhi- and HOXAlo-patients. HOXAlo-patients showed a reduced HOXA gene transcription, but instead overexpressed the homeobox gene IRX1. This transcriptional pattern was associated with a higher relapse rate and worse outcome. Here, we demonstrate that IRX1 binds to the MLL-AF4 complex at target gene promotors and counteract its promotor activating function. In addition, IRX1 induces transcription of HOXB4 and EGR family members. HOXB4 is usually a downstream target of c-KIT, WNT and TPO signaling pathways and necessary for maintaining and expanding in hematopoietic stem cells. EGR proteins control a p21-dependent quiescence program for hematopoietic stem cells. Both IRX1-dependend actions may help t(4;11) leukemia cells to establish a stem cell compartment. We also demonstrate that HDACi administration is functionally interfering with IRX1 and MLL-AF4, a finding which could help to improve new treatment options for t(4;11) patients.
The Asian tiger mosquito, Aedes albopictus (Diptera: Culicidae, SKUSE), is an important threat to public health due to its rapid spread and its potential as a vector. The eggs of Ae. albopictus are the most cold resistant life stage and thus, the cold hardiness of eggs is used to predict the future occurrence of the species in distribution models. However, the mechanism of cold hardiness has yet to be revealed. To address this question, we analyzed the layers of diapausing and cold acclimatized eggs of a temperate population of Ae. albopictus in a full factorial test design using transmission electron microscopy. We reviewed the hypotheses that a thickened wax layer or chorion is the cause of cold hardiness but found no evidence. As a result of the induced diapause, the thickness of the dark endochorion as a layer of high electron density and thus an assumed location for waxes was decreasing. We therefore hypothesized a qualitative alteration of the wax layer due to compaction. Cold acclimation was causing an increase in the thickness of the middle serosa cuticle indicating a detachment of serosa membrane from the endochorion as a potential adaptation strategy to isolate inoculating ice formations in the inter-membranous space.
This conference report comprises the contributions of European and American specialists in Fascism on the topic of networks, promises for the future and cultures of violence in Europe, 1922–1945. It was concluded that a much more in-depth examination of fascist networks, as well as their learning and acquisition processes is required, especially after 1939 and in the currently under-researched regions of Eastern and Southern-Eastern Europe. Secondly, the concept of a ‘New Man’ should be applied in more detailed studies on population and educational policies. Thirdly, there is a need to counter the frequently lamented asymmetrical state of research between Italian fascism and National Socialism.
Research in cell and developmental biology requires the application of three-dimensional model systems that reproduce the natural environment of cells. Processes in developmental biology are therefore studied in entire systems like insects or plants. In cell biology, three-dimensional cell cultures (e.g. spheroids or organoids) model the physiology and pathology of cells, tissues or organs. In all systems, the cellular neighborhood and interactions, but also physicochemical influences, are realistically presented. The production and handling of these model systems is rather simple and allows for reproducible characterization.
Confocal and light sheet-based fluorescence microscopy (LSFM) enable the observation of these systems while maintaining their three-dimensional integrity. LSFM is applicable to imaging live samples at high spatio-temporal resolution over long periods of time. The quality of the acquired datasets enables the extraction of quantitative features about morphology, functionality and dynamics in the context of the complete system. This approach is referred to as image-based systems biology. Exploiting the potential of the generated datasets requires an image analysis pipeline for data management, visualization and the retrieval of biologically meaningful values.
The goal of this thesis was to identify, develop and optimize modules of the image analysis pipeline. The modules cover data management and reduction, visualization, reconstruction of multiview image datasets, the segmentation and tracking of cell nuclei and the extraction of quantitative features. The modules were developed in an application-driven manner to test and ensure their applicability to real datasets from three-dimensional fluorescence microscopy. The underlying datasets were taken from research projects in developmental biology in insects and plants, as well as from cell biology.
The datasets acquired in fluorescence microscopy are typically complex and require common image processing steps in order to manage, visualize, and analyze the datasets. The first module accomplishes automatic structuring of large image datasets, reduces the data amount by image cropping and compression and computes maximum projection images along different spatial directions. The second module corrects for intensity variations in the generated maximum projection images that occur as a function of time. The program was published as a part of an article in Nature Protocols. Another developed module named BugCube provides a web-based platform to visualize and share the processed image datasets.
In LSFM, samples can be rotated in-between two acquisitions enabling the generation of multiview image datasets. Prior to my work, Frederic Strobl and Alexander Ross acquired the complete embryogenesis of the red flour beetle, Tribolium castaneum, and the field cricket, Gryllus bimaculatus, with LSFM. I evaluated a plugin for the software FIJI as a module for the reconstruction of such datasets. The plugin was optimized for automation and efficiency. We obtained the first high quality three-dimensional reconstructions of Tribolium and Gryllus datasets.
Optical clearing increases the penetration depth into samples, thus providing endpoint images of entire three-dimensional objects with cellular detail. This work contributes a quantitative characterization module that was applied to endpoint images of optically cleared spheroids. A program for the generation of ground truth datasets was developed in order to evaluate the cell nuclei segmentation performance. The program was part of a paper that was published in BMC Bioinformatics. Using the program, I could show that the cell nuclei segmentation is robust and accurate. Approaches from computational topology and graph theory complete the segmentation of cell nuclei. Thus, the developed module provides a comprehensive quantitative characterization of spheroids on the level of the individual cell, the cell neighborhood and the whole cell aggregate. The module was employed in four applications to analyze the influence of different stress conditions on the morphology and cellular arrangement of cells in spheroids. The module was accepted for publication in Scientific Reports along with the results for one application. The cell nuclei segmentation further provided a data source for simulation models that used correlation functions to identify structural zones in spheroids. These results were published in Royal Society Interface.
The final part of this work presents a module for cell tracking and lineage reconstruction. In collaboration with Dr. Alexis Maizel, Dr. Jens Fangerau and Dr. Daniel von Wangenheim, I developed a module to track the positions of all cells involved in lateral root formation in Arabidopsis thaliana and used the extracted positions for extensive data analysis. We reconstructed the cell lineages and established the first atlas of all founder cells that contribute to the formation. The analysis of the retrieved data allowed us to study conserved and individual patterns in lateral root formation. The atlas and parts of the analysis presented in this thesis were published in Current Biology.
In this thesis, I developed modules for an image analysis pipeline in three-dimensional fluorescence microscopy and applied them in interdisciplinary research projects. The modules enabled the organization, processing, visualization and analysis of the datasets. The perspective of the image analysis pipeline is not restricted to image-based systems biology. With ongoing development of the image analysis pipeline, it can also be a valuable tool for medical diagnostics or industrial high-throughput approaches.
The full-length translation-regulating add adenine riboswitch (Asw) from Vibrio vulnificus has a more complex conformational space than its isolated aptamer domain. In addition to the predicted apo (apoA) and holo conformation that feature the conserved three-way junctional purine riboswitch aptamer, it adopts a second apo (apoB) conformation with a fundamentally different secondary structure. Here, we characterized the ligand-dependent conformational dynamics of the full-length add Asw by NMR and by single-molecule FRET (smFRET) spectroscopy. Both methods revealed an adenine-induced secondary structure switch from the apoB-form to the apoA-form that involves no tertiary structural interactions between aptamer and expression platform. This strongly suggests that the add Asw triggers translation by capturing the apoA-form secondary structure in the holo state. Intriguingly, NMR indicated a homogenous, docked aptamer kissing loop fold for apoA and holo, while smFRET showed persistent aptamer kissing loop docking dynamics between comparably stable, undocked and docked substates of the apoA and the holo conformation. Unraveling the folding of large junctional riboswitches thus requires the integration of complementary solution structural techniques such as NMR and smFRET.

Es wird eine systematische Anleitung gegeben zur Revision der Namen von Assoziationen gemäß den Regeln des Internationalen Code der Pflanzensoziologischen Nomenklatur (ICPN, 3. Aufl. 2000). Außerdem werden die Verfahren zur gültigen Veröffentlichung von Assoziationen und Subassoziationen erläutert. Eine Übersicht über die Formen der Autorzitate und Zusätze bei syntaxonomischen Namen ist beigefügt.
Leonurus marrubiastrum ist eine hapaxanthe Art, die in der Regel zweijährig ist, unter günstigen Umständen ihren Lebenszyklus aber auch in einem Jahr vollenden kann. Unter natürlichen Bedingungen erreicht die Art leicht eine Wuchshöhe zwischen 1,2 m und 1,7 m. Die einsamigen Nüßchen können unter Wasser keimen und sich anschließend am Ufer etablieren. Die Art erweist sich im Experiment als nitrophil. Leonurus marrubiastrum zeigte in Mitteleuropa im vergangenen Jahrhundert einen starken Rückgang, insbesondere an Ruderalstandorten. Seit ca. 20 Jahren ist jedoch entlang der Elbe eine deutliche Zunahme der Populationsgrößen zu konstatieren. Ihren Schwerpunkt hat die Art in Deutschland derzeit in flussnahen Artemisietea-Gesellschaften, darüber hinaus an der Elbe auch im Chenopodion rubriund im Sisym brion. Als relativ kurzlebige Art ist Leonurus marrubiastrum auf gestörte Flächen angewiesen: Vorkommen im Chenopodion rubri in Nähe der Uferspülsäume sowie in tiefer gelegenen Buhnenfeldern, im Bereich des Mittelwassers auf Buhnen und Uferwerken, vor allem aber an höher gelegenen Uferabbrüchen und sonstigen Störstellen im Senecionion fluviatilis, zugleich oft im Halbschatten von Bäumen. Im südöstlichen Mitteleuropa ist Leonurus marrubiastrum auch Bestandteil dörflicher Ruderalfluren des Arction. Insgesamt kann Leonurus marrubiastrum daher nur als Artemisietea-Kennart eingestuft werden.
Endometriosis and its global research architecture : an in-depth density-equalizing mapping analysis

Background: Endometriosis is one of the most common gynecological diseases. It is still a chameleon in many aspects and urges intense research activities in the fields of diagnosis, therapy and prevention. Despite the need to foster research in this area, no in-depth analysis of the global architecture of endometriosis research exists yet.
Methods: We here used the NewQIS platform to conduct a density equalizing mapping study, using the Web of Science as database with endometriosis related entries between 1900 and 2009. Density equalizing maps of global endometriosis research encompassing country-specific publication activities, and semi-qualitative indices such as country specific citations, citation rates, h-Indices were created.
Results: In total, 11,056 entries related to endometriosis were found. The USA was leading the field with 3705 publications followed by the United Kingdom (952) and Japan (846). Concerning overall citations and country-specific h-Indices, the USA again was the leading nation with 74,592 citations and a modified h-Index of 103, followed by the UK with 15,175 citations (h-Index 57). Regarding the citation rate, Sweden and Belgium were at top positions with rates of 22.46 and 22.26, respectively. Concerning collaborative studies, there was a steep increase in numbers present; analysis of the chronological evolution indicated a strong increase in international collaborations in the past 10 years.
Conclusions: This study is the first analysis that illustrates the global endometriosis research architecture. It shows that endometriosis research is constantly gaining importance but also underlines the need for further efforts and investments to foster research and ultimately improve endometriosis management on a global scale.
Thought structures of modelling task solutions and their connection to the level of difficulty

Although efforts have been made to integrate the concept of mathematical modelling in school, among others PISA and TIMSS revealed weaknesses of not only German students in the field of mathematical modelling. There may be various reasons starting from educational policy via curricular issues to practical instructional concerns. Studies show that mathematical modelling has not been arrived yet in everyday school class (Blum &BorromeoFerri, 2009, p. 47). Thus, the proportion of mathematical modelling in everyday school classes is low (Jordan et al., 2006). When focusing on the teachers’ point of view there are difficulties which may contribute to avoid modelling tasks in class. The development of reasonable modelling tasks, estimating the task space, valuating the task difficulty and assessing the student solutions are difficulties which occur to an increasing degree compared to ordinary mathematics tasks.The project MokiMaS (transl.: modeling competency in math classes of secondary education) aims at providing inter-year modelling tasks, whose task space and level of difficulty is known, together with an evaluation scheme. In particular a theory based method has been developed to determine the level of difficulty of modelling tasks on the basis of thought structures, representing the cognitive load of solution approaches. The current question is whether this method leads to a realistic rating. To go further into that question an evaluation scheme has been developed which is guided by the daily assessment work of teachers, to investigate the relation of task difficulty and student performance.
SDF-1/CXCR4 expression in head and neck cancer and outcome after postoperative radiochemotherapy

Introduction: Outcome after postoperative radiochemotherapy (RT-CT) for patients with advanced head and neck squamous cell carcinomas (HNSCC) remains unsatisfactory, especially among those with HPV negative tumours. Therefore, new biomarkers are needed to further define subgroups for individualised therapeutic approaches. Preclinical and first clinical observations showed that the chemokine receptor CXCR4 and its ligand SDF-1 (CXCL12) play an important role in tumour cell proliferation, survival, cancer progression, metastasis and treatment resistance. However, the data on the prognostic value of SDF-1/CXCR4 expression for HNSCC are conflicting. The aim of our hypothesis-generating study was to retrospectively explore the prognostic potential of SDF-1/CXCR4 in a well-defined cohort of HNSCC patients collected within the multicenter biomarker study of the German Cancer Consortium Radiation Oncology Group (DKTK-ROG).
Material and methods: Patients with stage III and IVA HNSCC of the oral cavity, oropharynx and hypopharynx were treated with resection and adjuvant radiotherapy (RT) with ≥60 Gy and concurrent cisplatin-based chemotherapy (CT). Tissue micro-arrays (TMAs) from a total of 221 patients were generated from surgical specimens, 201 evaluated for the SDF-1 and CXCR4 expression by immunofluorescence and correlated with clinico-pathological and outcome data.
Results: In univariate and multivariate analyses intracellular SDF-1 expression was associated with lower loco-regional control (LRC) in the entire patient group as well as in the HPV16 DNA negative subgroup. CXCR4 expression showed a trend for lower LRC in the univariate analysis which was not confirmed in the multivariate analysis. Neither for SDF-1 nor CXCR4 expression associations with distant metastasis free or overall survival were found.
Conclusions: Our exploratory data support the hypothesis that overexpression of intracellular SDF-1 is an independent negative prognostic biomarker for LRC after postoperative RT-CT in high-risk HNSCC. Prospective validation is warranted and further exploration of SDF-1/CXCR4 as a potential therapeutic target to overcome treatment resistance in HNSCC appears promising.
Motivated by the necessary replacement of the GSI UNILAC poststripper linac, a compact and efficient linac design based on IH-type cavities has been developed. Using KONUS beam dynamics, it was possible to design a linac consisting of only five cavities that can be operated by the existing UNILAC RF amplifier structure. The transversal focusing scheme is based on magnetic quadrupole triplet lenses. The optimized design provides full transmission and low emittance growth for the design current of 15 emA U28+, accelerating the beam from 1.4 MeV/u to 11.4 MeV/u. Extensive error studies were performed to define tolerances and verify the stability of the design with respect to misalignment and injection parameters. The design provides a compact and cost effective alternative to a new Alvarez linac. With a total length of just 22.8 meters it will leave room for future energy upgrades in the UNILAC tunnel.
The Earth's future depends on how we manage the manifold risks of climate change (CC). It is state-of-the-art to assume that risk reduction requires participatory management involving a broad range of stakeholders and scientists. However, there is still little knowledge about the optimal design of participatory climate change risk management processes (PRMPs), in particular with respect to considering the multitude of substantial uncertainties that are relevant for PRMPs. To support the many local to regional PRMPs that are necessary for a successful global-scale reduction of CC risks, we present a roadmap for designing such transdisciplinary knowledge integration processes. The roadmap suggests ways in which uncertainties can be comprehensively addressed within a PRMP. We discuss the concept of CC risks and their management and propose an uncertainty framework that distinguishes epistemic, ontological, and linguistic uncertainty as well as ambiguity. Uncertainties relevant for CC risk management are identified. Communicative and modeling methods that support social learning as well as the development of risk management strategies are proposed for each of six phases of a PRMP. Finally, we recommend how to evaluate PRMPs as such evaluations and their publication are paramount for achieving a reduction of CC risks.
Allostery is a phenomenon observed in many proteins where binding of a macromolecular partner or a small-molecule ligand at one location leads to specific perturbations at a site not in direct contact with the region where the binding occurs. The list of proteins under allosteric regulation includes AGC protein kinases. AGC kinases have a conserved allosteric site, the phosphoinositide-dependent protein kinase 1 (PDK1)-interacting fragment (PIF) pocket, which regulates protein ATP-binding, activity, and interaction with substrates. In this study, we identify small molecules that bind to the ATP-binding site and affect the PIF pocket of AGC kinase family members, PDK1 and Aurora kinase. We describe the mechanistic details and show that although PDK1 and Aurora kinase inhibitors bind to the conserved ATP-binding site, they differentially modulate physiological interactions at the PIF-pocket site. Our work outlines a strategy for developing bidirectional small-molecule allosteric modulators of protein kinases and other signaling proteins.
In the cochlea of the mustached bat, cochlear resonance produces extremely sharp frequency tuning to the dominant frequency of the echolocation calls, around 61 kHz. Such high frequency resolution in the cochlea is accomplished at the expense of losing temporal resolution because of cochlear ringing, an effect that is observable not only in the cochlea but also in the cochlear nucleus. In the midbrain, the duration of sounds is thought to be analyzed by duration-tuned neurons, which are selective to both stimulus duration and frequency. We recorded from 57 DTNs in the auditory midbrain of the mustached bat to assess if a spectral-temporal trade-off is present. Such spectral-temporal trade-off is known to occur as sharp tuning in the frequency domain which results in poorer resolution in the time domain, and vice versa. We found that a specialized sub-population of midbrain DTNs tuned to the bat’s mechanical cochlear resonance frequency escape the cochlear spectral-temporal trade-off. We also show evidence that points towards an underlying neuronal inhibition that appears to be specific only at the resonance frequency.
Precise temporal coding is necessary for proper acoustic analysis. However, at cortical level, forward suppression appears to limit the ability of neurons to extract temporal information from natural sound sequences. Here we studied how temporal processing can be maintained in the bats’ cortex in the presence of suppression evoked by natural echolocation streams that are relevant to the bats’ behavior. We show that cortical neurons tuned to target-distance actually profit from forward suppression induced by natural echolocation sequences. These neurons can more precisely extract target distance information when they are stimulated with natural echolocation sequences than during stimulation with isolated call-echo pairs. We conclude that forward suppression does for time domain tuning what lateral inhibition does for selectivity forms such as auditory frequency tuning and visual orientation tuning. When talking about cortical processing, suppression should be seen as a mechanistic tool rather than a limiting element.
Background: Differential RNA-Seq (dRNA-Seq) is a recently developed method of performing primary transcriptome analyses that allows for the genome-wide mapping of transcriptional start sites (TSSs) and the identification of novel transcripts. Although the transcriptomes of diverse bacterial species have been characterized by dRNA-Seq, the transcriptome analysis of archaeal species is still rather limited. Therefore, we used dRNA-Seq to characterize the primary transcriptome of the model archaeon Haloferax volcanii.
Results: Three independent cultures of Hfx. volcanii grown under optimal conditions to the mid-exponential growth phase were used to determine the primary transcriptome and map the 5′-ends of the transcripts. In total, 4749 potential TSSs were detected. A position weight matrix (PWM) was derived for the promoter predictions, and the results showed that 64 % of the TSSs were preceded by stringent or relaxed basal promoters. Of the identified TSSs, 1851 belonged to protein-coding genes. Thus, fewer than half (46 %) of the 4040 protein-coding genes were expressed under optimal growth conditions. Seventy-two percent of all protein-coding transcripts were leaderless, which emphasized that this pathway is the major pathway for translation initiation in haloarchaea. A total of 2898 of the TSSs belonged to potential non-coding RNAs, which accounted for an unexpectedly high fraction (61 %) of all transcripts. Most of the non-coding TSSs had not been previously described (2792) and represented novel sequences (59 % of all TSSs). A large fraction of the potential novel non-coding transcripts were cis-antisense RNAs (1244 aTSSs). A strong negative correlation between the levels of antisense transcripts and cognate sense mRNAs was found, which suggested that the negative regulation of gene expression via antisense RNAs may play an important role in haloarchaea. The other types of novel non-coding transcripts corresponded to internal transcripts overlapping with mRNAs (1153 iTSSs) and intergenic small RNA (sRNA) candidates (395 TSSs).
Conclusion: This study provides a comprehensive map of the primary transcriptome of Hfx. volcanii grown under optimal conditions. Fewer than half of all protein-coding genes have been transcribed under these conditions. Unexpectedly, more than half of the detected TSSs belonged to several classes of non-coding RNAs. Thus, RNA-based regulation appears to play a more important role in haloarchaea than previously anticipated.
Background: As a multi-targeted anti-angiogenic receptor tyrosine kinase (RTK) inhibitor sunitinib (SUN) has been established for renal cancer and gastrointestinal stromal tumors. In advanced refractory esophagogastric cancer patients, monotherapy with SUN was associated with good tolerability but limited tumor response.
Methods: This double-blind, placebo-controlled, multicenter, phase II clinical trial was conducted to evaluate the efficacy, safety and tolerability of SUN as an adjunct to second and third-line FOLFIRI (NCT01020630). Patients were randomized to receive 6-week cycles including FOLFIRI plus sodium folinate (Na-FOLFIRI) once every two weeks and SUN or placebo (PL) continuously for four weeks followed by a 2-week rest period. The primary study endpoint was progression-free survival (PFS). Preplanned serum analyses of VEGF-A, VEGF-D, VEGFR2 and SDF-1α were performed retrospectively.
Results: Overall, 91 patients were randomized, 45 in each group (one patient withdrew). The main grade ≥3 AEs were neutropenia and leucopenia, observed in 56 %/20 % and 27 %/16 % for FOLFIRI + SUN/FOLFIRI + PL, respectively. Median PFS was similar, 3.5 vs. 3.3 months (hazard ratio (HR) 1.11, 95 % CI 0.70–1.74, P = 0.66) for FOLFIRI + SUN vs. FOLFIRI + PL, respectively. For FOLFIRI + SUN, a trend towards longer median overall survival (OS) compared with placebo was observed (10.4 vs. 8.9 months, HR 0.82, 95 % CI 0.50–1.34, one-sided P = 0.21). In subgroup serum analyses, significant changes in VEGF-A (P = 0.017), VEGFR2 (P = 0.012) and VEGF-D (P < 0.001) serum levels were observed.
Conclusions: Although sunitinib combined with FOLFIRI did not improve PFS and response in chemotherapy-resistant gastric cancer, a trend towards better OS was observed. Further biomarker-driven studies with other anti-angiogenic RTK inhibitors are warranted.
Trial registration: This study was registered prospectively in the NCT Clinical Trials Registry (ClinicalTrials.gov) under NCT01020630 on November 23, 2009 after approval by the leading ethics committee of the Medical Association of Rhineland-Palatinate, Mainz, in coordination with the participating ethics committees (see Additional file 2) on September 16, 2009.
In patients with glioblastoma, antiangiogenic therapy with bevacizumab (BEV) has been shown to improve progression-free survival (PFS), but not overall survival (OS). Especially in patients with an unusual infiltrative phenotype as seen in multifocal glioblastoma, the use of BEV therapy is still more controversial. Therefore, we prepared a retrospective case series with 16 patients suffering from a multifocal glioblastoma treated with BEV. We compared these patients to a matched control cohort of 16 patients suffering from glioblastoma with a single lesion treated with BEV. The objective of this study was to evaluate whether the course of disease differs in glioblastoma patients with a multifocal disease pattern compared to those with a single lesion only. Patients were treated with BEV monotherapy or BEV in combination with irinotecan or lomustine (CCNU). Response rates and PFS were similar in both groups. There was a trend for an unfavorable OS in the patient group with multifocal glioblastoma, which was expected due to the generally worse prognosis of multifocal glioblastoma. We investigated whether BEV therapy affects the invasive growth pattern as measured by the appearance of new lesions on magnetic resonance imaging (MRI). Under BEV therapy, there was a trend for a lower frequency of new lesions both in multifocal and solitary glioblastoma. Based on these results, BEV therapy at relapse appears to be justified to no lesser extent in multifocal glioblastoma than in solitary glioblastoma.
This article describes the motion database for a large sample (n = 2400) of 7-m penalty throws in team handball that includes 1600 disguised throws. Throws were performed by both novice (n = 5) and expert (n = 5) penalty takers. The article reports the methods and materials used to capture the motion data. The database itself is accessible for download via JLU Web Server and provides all raw files in a three-dimensional motion data format (.c3d). Additional information is given on the marker placement of the penalty taker, goalkeeper, and ball together with details on the skill level and/or playing history of the expert group. The database was first used by Helm et al. (2017) to investigate the kinematic patterns of disguised movements. Results of this analysis are reported and discussed in their article "Kinematic patterns underlying disguised movements: Spatial and temporal dissimilarity compared to genuine movement patterns" (doi:10.1016/j.humov.2017.05.010).
Cognitive flexibility, the ability to flexibly switch between tasks, is a core dimension of executive functions (EFs) allowing to control actions and to adapt flexibly to changing environments. It supports the management of multiple tasks, the development of novel, adaptive behavior and is associated with various life outcomes. Cognitive flexibility develops rapidly in preschool and continuously increases well into adolescence, mirroring the growth of neural networks involving the prefrontal cortex. Over the past decade, there has been increasing interest in interventions designed to improve cognitive flexibility in children in order to support the many developmental outcomes associated with cognitive flexibility. This article provides a brief review of the development and plasticity of cognitive flexibility across early and middle childhood (i.e., from preschool to elementary school age). Focusing on interventions designed to improve cognitive flexibility in typically developing children, we report evidence for significant training and transfer effects while acknowledging that current findings on transfer are heterogeneous. Finally, we introduce metacognitive training as a promising new approach to promote cognitive flexibility and to support transfer of training.
Anthropogenic changes in climate and land use are driving changes in migration patterns of birds worldwide. Spatial changes in migration have been related to long-term temperature trends, but the intrinsic mechanisms by which migratory species adapt to environmental change remain largely unexplored. We show that, for a long-lived social species, older birds with more experience are critical for innovating new migration behaviours. Groups containing older, more experienced individuals establish new overwintering sites closer to the breeding grounds, leading to a rapid population-level shift in migration patterns. Furthermore, these new overwintering sites are in areas where changes in climate have increased temperatures and where food availability from agriculture is high, creating favourable conditions for overwintering. Our results reveal that the age structure of populations is critical for the behavioural mechanisms that allow species to adapt to global change, particularly for long-lived animals, where changes in behaviour can occur faster than evolution.
Whiteout: animal traces in Werner Herzog’s Grizzly man and encounters at the end of the world

Literary animal studies are confronted with a systematic question: How can writing, as a human-made sign system, represent the nonhuman animal as an autonomous agent without falling back into the pitfalls of anthropomorphism? Against the backdrop of this problem, this paper asks how the medium of film allows for a different representation of the animal and analyzes two of Werner Herzog’s later documentary films. Although the depiction of animals and landscapes has always played a significant part in Herzog’s films, critical assessments of his work—including those of Herzog himself—tended to view the role of nature imagery as purely allegorical: it expresses the inner nature, the inner landscapes of the film’s human protagonists. This paper tries to open up a different view. It argues that both Grizzly Man and Encounters at the End of the World develop an aesthetic that depicts nonhuman nature as an autonomous and lively presence. In the close proximity amongst camera, human, and nonhuman agents, a clear distinction between nature and culture is increasingly blurred.
Relativistic jets from active galactic nuclei (AGN) often display a non-uniform structure and are, under certain conditions, susceptible to a number of instabilities. An interesting example is the development of non-axisymmetric, Rayleigh-Taylor type instabilities in the case of differentially rotating two-component jets, with the toroidal component of the magnetic field playing a key role in the development or suppression of these instabilities. We have shown that higher magnetization leads to stability against these non-axisymmetric instabilities. Using ray-casting on data from relativistic MHD simulations of two-component jets, we now investigate the effect of these instabilities on the synchrotron emission pattern from the jets. We recover many well known trends from actual observations, e.g., regarding the polarization fraction and the distribution of the position angle of the electric field, in addition to a different emitting region, depending on the stability of the jet.
Die Wissens'poetologie' bezeichnet eine 'Praxis', die ein Tableau verschiedener Wissensfelder freilegt, um die Bedingungen der historisch-epistemologischen Möglichkeiten verschiedener Wissensordnungen zu hinterfragen und zu untersuchen. Doch wie werden diese Praktiken 'in concreto' von der Literatur vollzogen? Zwei mögliche wissenspoetologische Praktiken sollen anhand der Schriften Walter Benjamins und Jorge Luis Borges' verdeutlicht werden, um sie weiterführend als Paradigma für komparatistische Nachforschungen in der Wissenspoetologie zu verwenden: die 'Fiktionalisierung' und das 'Experiment'.
Das Wechselspiel von kulturellen und hermeneutischen Grenzerfahrungen bildet die wohl deutlichste Gemeinsamkeit zwischen Borges und Jarmusch: Beide Autoren verorten ihre Figuren in zahlreichen Erzählungen an der Schnittstelle verschiedener und bisweilen gegensätzlicher Kulturen und Lebenswelten, die sich im Inneren der Figur gegenseitig durchdringen. Während Borges vor allem in seinem Frühwerk kulturelle Grenzsituationen entwirft, die den Gegensatz zwischen der europäischen und kreolischen Kultur zum Ausdruck bringen, widmen sich Jarmuschs Filme insbesondere den kulturellen Spannungen in den Vereinigten Staaten. Doch gleichzeitig entwerfen sie die Utopie einer 'Hyperkultur', in der die Gegensätze zugunsten kultureller Synergien aufgehoben sind. Als kulturelle Grenzgänger oszillieren ihre Figuren auf einer Schwelle zwischen den Kulturen, die sich den agonal-dialektischen Gegensätzen kontinuierlich entzieht (vgl. Han 2005, 17). Gleichzeitig legen sie dem Phänomen der kulturellen Heterogenität eine Erzählstruktur zugrunde, die selbst von einer wesentlichen Ambiguität geprägt ist und die Dynamik kultureller Grenzerfahrungen überhaupt erst sichtbar macht. Borges' berühmte Erzählung 'El Sur' und Jarmuschs Kunstfilm 'Dead Man' offenbaren dabei erstaunliche strukturelle Gemeinsamkeiten.

Daß in die Visionen des Jorge Luis Borges Reminiszenzen an Werke der bildenden Kunst eingeflossen sind, erscheint evident, wenn man etwa die Beschreibung der 'La biblioteca de Babel' (Borges 1989a, 465-471) mit den Kerkerphantasien Giovanni Battista Piranesis ('Carceri', 1745-1750) oder auch mit der Darstellung des Babylonischen Turmbaus (1563) durch Pieter Breughel d.Ä. vergleicht. Auch bestehen vielfaltige Beziehungen der Borges'schen Labyrinth-, Spiegel- und Doppelgängerphantasien zu Gemälden der Surrealisten sowie zu anderen Werken, insbesondere der modernen Kunst. Inzwischen hat Borges der bildenden Kunst gleichsam mit Zinsen zurückerstattet, was er ihr verdankt. Bildende Künstler verschiedener Stilrichtungen haben aus seinen Werken Anregungen bezogen, Borges'sche Visionen visualisiert, mit Gemälden, Photographien und Graphiken auf die Denkbilder des Argentiniers geantwortet.
Die Parallelen zwischen Cortázars 'Rayuela' und Musils 'Mann ohne Eigenschaften' wurden von wissenschaftlicher Seite schon mehrfach untersucht. Die Frage, ob und inwieweit sich die Berührungspunkte der Romane auf einen direkten Einfluss Musils zurückführen lassen, ist dabei weitgehend ausgeklammert geblieben. Einen ersten Schritt dahingehend soll hier durch die Aufarbeitung von Umständen und Umfang von Cortázars Musil-Rezeption, die bisher ebenso wenig beachtet wurde, getan werden. Ein weiterer Schritt ist die Analyse von Kontext und Funktion, die den Nennungen Musils in 'Rayuela' zukommt. Die im Zuge der Rezeptionsaufarbeitung durch die Durchsicht von Cortázars Exemplaren der Musilschen Werke gewonnenen Kenntnisse über jene Passagen, die ihm bedeutend erschienen sein dürften, erleichtern es außerdem, Musilsche Intertexte in 'Rayuela' aufzuspüren, die als Indiz eines Einflusses gewertet werden können. Dass es davon mehr gibt, als bisher angenommen, soll an einer exemplarischen Analyse des 71. Kapitels von 'Rayuela' verdeutlicht werden.
Ausgehend von zwei Punkten - die Gefühle als zentraler Inhalt dieser Werke und ihre spezifische aus dem Sujet hervorgehende Form - lassen sich Gemeinsamkeiten zwischen Ovid und Kluge beobachten, die die Substanz dieser Werke betreffen. Das bedeutet, wenn wir zunächst bei der Form bleiben, dass Ovids Werk keine Ganzheit ist, jedenfalls keine Ganzheit im traditionellen Sinn, und das bedeutet umgekehrt, dass Kluges Werk die 'disiecta membra' eines Ganzen entwirft, auf das es sich virtuell zubewegt. Die These ist, dass Kluges und Ovids Werk von derselben Formidee getragen ist und dass diese sich dieser Formidee lediglich von zwei verschiedenen Seiten nähern, Kluge von der Seite der modernen Kleinstform des Fragments, Ovid von der Seite der antiken Großform des Epos.
In dieser Betrachtung wird versucht, die Besonderheiten und Schwächen von Ehrensteins 'Pe-Lo-Thien' zu erläutern und aus einer chinesischen Perspektive zu bewerten, ob seine Nachdichtungen den ursprünglichen Stil und Wert von Pe Lo Thiens Gedichten beibehalten, ferner: durch die Analyse der Hauptthemen und des Sprachgebrauchs herauszufinden, ob seine Nachdichtungen expressionistische Elemente enthalten. Außerdem wird Pe Lo Thiens Gedicht 'In der Nacht einen Singenden hören. Übernachten in E Zhou' mit Ehrensteins Nachdichtung 'Ein Mensch weint in der Nacht' und mit Klabunds Nachdichtung 'Das nächtliche Lied und die fremde Frau' verglichen, um die Mehrdeutigkeit von Pe Lo Thiens Gedicht aufzuzeigen und einige Unterschiede zwischen der westlichen und der chinesischen Dichtung zu verdeutlichen.
Meine Ausführungen gliedern sich in zwei Teile. Zunächst zeige ich anhand einer Analyse der Charaktere der Erzählung 'Der Bruder des Gavillans', wie Bodo Uhse sich in die zeitgenössische Diskussion um die mexikanische Identität einfugt, indem er seine Figuren durch Rückgriff auf wesentliche Kennzeichen des 'homo mexicanus' gestaltet. Im Anschluss werden die gewonnenen Ergebnisse anhand der Erzählung 'Reise in einem blauen Schwan' überprüft und erweitert.
Die Erfahrung des Exils spielt im Leben Iosif Brodskijs eine große Rolle, zumal er nach seiner Umsiedlung nach Amerika wusste, dass er niemals nach Russland würde zurückkehren können und sein weiteres Schaffen als Schriftsteller in einer neuen, fremdsprachigen Umgebung würde gestalten müssen. Bevor ich auf die hier im Vordergrund stehende exilbedingte Sprachproblematik bei Brodskij eingehe, möchte ich einige allgemeine Anmerkungen zum Exil vorwegschicken, damit man Brodskijs Verarbeitungen der Exilereignisse und die damit zusammenhängenden Auseinandersetzungen in seinen Texten besser verstehen kann.
Der römische Kaiser Claudius, dritter Prinzeps nach Augustus, regierte von 41 – 54 n. Chr. und wurde vermutlich von Agrippina, seiner letzten Ehefrau, mit einem Pilzgericht vergiftet. Obwohl er wegen seiner angeborenen Körperbehinderung als dynastischer Nachfolger nicht vorgesehen war, folgte er C. Caligula unmittelbar nach dessen Ermordung auf den Kaiserthron.
Die dem Amt inhärenten strukturellen Schwierigkeiten wurden verstärkt durch seine imbecillitas, die nicht nur Seneca, seinen Zeitgenossen, sondern auch Tacitus, Sueton und Cassius Dio, die ihn nachfolgend zum Sujet ihrer Werke machen, erheblich verunsicherte.
Besonders die Vertreter der antiken Historiographie und Biographie stehen vor der Herausforderung, glaubhaft erklären zu müssen, warum ein imbecillus, der Gegenentwurf zu Augustus, an die Macht gelangen und sie 13 Jahre behalten konnte, ohne unterschiedlich motivierten Anschlägen zum Opfer zu fallen. Aus dieser Diskrepanz entstehen – abhängig von der persönlichen Vorstellung des jeweiligen Autors und den gesellschaftlich bedingten Vorurteilen über Behinderung – unterschiedliche Bilder, die nicht nur das Dilemma des behinderten Kaisers, sondern auch das seiner Interpreten illustrieren: Claudius tritt, je nach Situation und Interaktionspartnern, als willenlos Re-agierender oder als taktisch Agierender auf: Als Regierenden hingegen zeigen ihn die Berichte gar nicht oder selten. Unberechenbarkeit, Übertreibung und Wiederholung werden für die antiken Schriftsteller zum Herrschafts- und Persönlichkeitsmerkmal des Prinzeps, die Antithese zum Narrativ.
Das essentielle Kommunikationsproblem des behinderten Claudius scheint, folgt man den Berichten, aus einem bewussten oder charakterbedingten Verzicht auf die patria potestas zu resultieren, mit katastrophalen Auswirkungen auf sein Ansehen als Prinzeps. Die von allen Autoren kritisierte Abhängigkeit von den Mitgliedern seines Hofes, zu deren prominentesten Opfern die junge und unerfahrene Messalina und letztlich Claudius selbst zählen, erregt Spott und Unmut der Plebs und verunsichert die Aristokratie durch eine als willkürlich empfundene Rechtsprechung.
Andererseits verweisen die Darstellungen seiner erstaunlichen Amtserhebung, des siegreichen Britannienfeldzugs, des abrupten Sturzes Messalinas, aber auch Agrippinas plötzlicher Angst, die sie zum Gattenmord veranlasst, auf einen Kaiser, der bei Bedarf offenbar recht gezielt das Klischee des imbecillus als Mittel des Machterhalts zu bedienen vermag.
So entsteht aus Claudius, abhängig vom gesellschaftlichen Status Behinderter, in den antiken und modernen Schriften, die sich mit ihm und seiner Amtsführung befassen, die paradoxe Figur eines schuldigen Opfers bzw. unschuldigen Täters: Sowohl Opfer- als auch Täterrolle sind a priori durch die Behinderung gerechtfertigt und relativiert.
Aufbauend auf den Erfahrungen zweier Workshops zu (urbaner) Austerität in Griechenland und Deutschland diskutiert der Beitrag die (unterschiedliche) Geschichte und Geographie der Austerität mit besonderem Blick auf die Regionen Frankfurt/Rhein-Main und Athen. Die Erfahrungen der multiplen Krise seit 2008, die sich in Griechenland vor dem Hintergrund einer austeritätspolitischen "Shock Doctrine" und in der BRD im Kontext eines langfristigen Projekts der "scheibchenweisen" Austerität entwickelten, eröffnen dabei die Möglichkeit, die Debatten um urbane Austerität einem kritischen Blick zu unterziehen. Der Beitrag sieht insbesondere im Bereich der Krisen der (urbanen) sozialen Reproduktion sowie der Krisen der (städtischen) Politik und Repräsentation weiteren Forschungsbedarf.
Background: Lectures remain an important teaching method to present and structure knowledge to many students concurrently. Adequate measures are necessary to maintain the quality of the lectures. The aim of this study was to determine the impact on the lecture quality using written structured feedback and to compare the ratings of surgical lectures between students and surgical peers.
Methods: Prospective analysis of two consecutive surgical lecture series for undergraduate students at Goethe-University Medical School was performed before and after evaluation of the lecturers via independent written feedback from trained undergraduate students and surgeons. The 22-item feedback instrument covered three areas of performance: content, visualization, and delivery. Additional suggestions for improvement were provided from
both students and surgical peers who anonymously attended the lectures. The lecturers, experienced surgeons, as well as the student and peer raters were blinded in terms of the aim and content of the study. Their response to the feedback was collected using a web-based 13-item questionnaire. The Kendall’s-W coefficient was computed to calculate inter-rater reliability (IRR). Differences between ratings before and after feedback were analyzed using Student’s t-test for dependent samples. The Kolmogorov-Smirnov-test was used for independent samples.
Results: A total of 22 lectures from a possible 32 given by 13 lecturers were included and analyzed by at least three surgeons and two students. There were significant improvements in overall score as well as in the details of 9 of the 13 items were found. The average inter-rater reliability was 0.71. There were no differences in the ratings as a function of the rater’s level of expertise (peers vs. students). We found that 13/23 lecturers (56.5%) answered the questionnaire, and 92% strongly agreed that the written feedback was useful. 76.9% of the lecturers revised their lecture based on the written feedback requiring on average 112.5 min (range from 20 to 300 min).
Conclusions: Overall, this study indicates that structured written feedback provided by trained peers and students that is subsequently discussed by the lecturers concerned is a highly effective and efficient method to improve aspects of lecturing. We anticipate that structured written feedback by trained students that is discussed by the lecturers concerned will improve lecturing.

General intelligence is a psychological construct that captures in a single metric the overall level of behavioural and cognitive performance in an individual. While previous research has attempted to localise intelligence in circumscribed brain regions, more recent work focuses on functional interactions between regions. However, even though brain networks are characterised by substantial modularity, it is unclear whether and how the brain’s modular organisation is associated with general intelligence. Modelling subject-specific brain network graphs from functional MRI resting-state data (N = 309), we found that intelligence was not associated with global modularity features (e.g., number or size of modules) or the whole-brain proportions of different node types (e.g., connector hubs or provincial hubs). In contrast, we observed characteristic associations between intelligence and node-specific measures of within- and between-module connectivity, particularly in frontal and parietal brain regions that have previously been linked to intelligence. We propose that the connectivity profile of these regions may shape intelligence-relevant aspects of information processing. Our data demonstrate that not only region-specific differences in brain structure and function, but also the network-topological embedding of fronto-parietal as well as other cortical and subcortical brain regions is related to individual differences in higher cognitive abilities, i.e., intelligence.
In der vorliegenden Arbeit wurde die Dynamik zweier grundlegend verschiedener, deaktivierender Mechanismen von Retinalproteinen untersucht. In einem dritten Projekt wurde die Photodynamik einer Dreifachmutante von visuellem Rhodopsin erforscht, von der eine Mutation zu kongenitaler (angeborener) Nachtblindheit führt und zwei andere Mutationen das Protein über eine Disulfidbrücke stabilisieren. Die Ergebnisse dieser drei Projekte sind im Folgenden zusammengefasst.
Die Aktivität des mikrobiellen Proteorhodopsins als lichtgetriebene Protonenpumpe kann photoinduziert unterbunden werden. Dies erfolgt durch die Absorption von blauem Licht durch das Retinal bei deprotonierter Schiff‘schen Base. Vor dieser Arbeit war allerdings nur wenig über den Mechanismus und die Kinetik dieses Effekts bekannt. Das einzige Retinalprotein, an dem diese Deaktivierungsdynamik auf molekularer Ebene zeitaufgelöst untersucht wurde, ist Bakteriorhodopsin. Doch auch an diesem System wurde die ultraschnelle Primärreaktion in der photoinduzierten Deaktivierungsdynamik - die Photoisomerisierung des 13-cis-Retinals - bisher nicht zeitaufgelöst gemessen.
In dieser Arbeit wurde ein Weg gefunden, diesen Prozess auf einer Sub-Pikosekundenzeitskala zu detektieren. Dazu wurde eine Proteorhodopsinmutante genutzt, in der der primäre Protonendonor E108 durch Glutamin ersetzt ist. Diese Mutante weist eine signifikante Erhöhung der Lebensdauer des M-Intermediats auf. Im photostationären Gleichgewicht führt diese veränderte Kinetik zu einer erheblich erhöhten Akkumulation des Proteins im M-Zustand, die ausreicht, um photoinduzierte Absorptionsänderungen der Deaktivierungsdynamik sowohl im sichtbaren als auch im mittleren Infrarotbereich auf ultrakurzer Zeitskala zu detektieren. Dieses Projekt erfolgte in Kooperation mit dem Arbeitskreis Glaubitz (Goethe-Universität Frankfurt am Main).
Es zeigte sich, dass die Anregung des Retinals von Proteorhodopsin im M-Zustand zur Isomerisierung von 13-cis zu all-trans führt, die nach wenigen Pikosekunden abgeschlossen ist. Der zweite und abschließende Schritt ist die Reprotonierung der Schiff'schen Base. Es stellte sich heraus, dass dieser Prozess auf einer Nanosekundenzeitskala abläuft und über einen Protonentransfer vom primären Protonenakzeptor D97 zur Schiff'schen Base ermöglicht ist.
Die in dieser Arbeit vorgestellte Methodik zur Untersuchung der deaktivierenden Photodynamik von Proteorhodopsin auf ultraschneller Zeitskala, könnte in Zukunft auf weitere mikrobielle Rhodopsine angewandt werden. So ist die Studie der Deaktivierungsdynamik von Channelrhodopsinen von großem Interesse für optogenetische Anwendungen. Eine lichtgesteuerte Kontrolle der Ionenkanalöffnung und -schließung sollte die Präzision in der Regulierung ionischer Permeation erheblich verbessern.
Die Proteorhodopsinmutante E108Q wurde außerdem in ihrer primären Photodynamik sowohl bei grünem als auch blauem Anregungslicht untersucht. Es zeigte sich in beiden Fällen eine Dynamik, die der des Wildtyps sehr ähnlich ist. Eine Beobachtung unterscheidet sich jedoch wesentlich vom Wildtyp. Das K-Intermediat der E108Q-Mutante scheint nach einigen hundert Pikosekunden zumindest partiell zu zerfallen, woraufhin sich eine Signatur im blauen Spektralbereich bildet. Blitzlichtphotolysemessungen lassen vermuten, dass diese blau absorbierende Species im zwei- bis dreistelligen Nanosekundenbereich wieder zerfallen sein muss.
Der zweite Teil dieser Arbeit beschäftigt sich mit dem Photozerfall von visuellem Rhodopsin. Es ist bekannt, dass die Signaltransduktion durch Wechselwirkung zwischen aktiviertem Rhodopsin und Arrestin unterbunden wird. Im ersten Abschnitt wurde der Einfluss der Arrestin-1-Variante p44 auf die Photodynamik visuellen, bovinen Rhodopsins untersucht. In einer Kooperation mit dem Arbeitskreis Schwalbe (Goethe-Universität Frankfurt am Main) konnte gezeigt werden, dass Arrestin erheblichen Einfluss auf die Zerfallsdynamik von Meta II und Meta III hat. Es wurde festgestellt, dass die Wechselwirkung von p44 mit photoaktiviertem Rhodopsin eine erhöhte Population des Intermediats Meta III bewirkt, mit der Folge einer zweifach langsameren Freisetzungskinetik des all-trans-Retinals. Diese Beobachtung weist auf eine physiologische Rolle des Zustands Meta III in der Retinalhomöostase hin.
Gegenstand einer zweiten Studie mit dem Arbeitskreis Schwalbe ist zum einen die Rhodopsinmutation G90D, die mit kongenitaler (angeborener) stationärer Nachtblindheit zusammenhängt, und zum anderen die Doppelmutation N2C und D282C, die zur Ausbildung einer stabilisierenden Disulfidbrücke zwischen den im extrazellulären Bereich eingeführten Cysteinen führt. Im Rahmen dieser Arbeit wurde die Photodynamik des Wildtyps, der Doppelmutante und der stabilisierten G90D-Mutante (Mutationen G90D, N2C und D282C) sowohl auf einer ultrakurzen Zeitskala als auch auf einer Minutenskala untersucht.
Die vorliegende Arbeit untersucht die Möglichkeiten Sozialer Arbeit in segregierten städtischen Gebieten unter den Bedingungen der Festivalisierung von Stadtentwicklungspolitik. Der Hamburger Stadtteil Wilhelmsburg, bisher vorrangig als „benachteiligtes“ oder „gefährliches“ Gebiet charakterisiert, stand bis zum Jahr 2013 im Mittelpunkt des Rahmenprogramms Sprung über die Elbe, dessen zentrale Maßnahmen eine „Internationale Bauausstellung 2013 (IBA)“ und eine „internationale gartenschau 2013 (igs)“ waren. Wenn ein solches Gebiet mit dem Repertoire der unternehmerischen Stadtpolitik entwickelt wird, verändert sich das Verhältnis von Sozialer Arbeit und Stadtentwicklungspolitik.
Eine zentrale Fragestellung der Arbeit beschäftigt sich mit der Veränderung der institutionellen Landschaft in Stadtentwicklungsprozessen: Welche Ausgangslage findet Soziale Arbeit in segregierten Gebieten unter den Bedingungen einer neoliberalisierten Stadtpolitik vor? Ein weiterer Schwerpunkt liegt auf der Untersuchung der Zugänglichkeit von Ressourcen, die die Bewohner_innen des Gebiets für ihre räumliche Reproduktion einsetzen können.
Auffällig am Vorgehen der Hamburger Stadtentwicklungspolitik im Sprung über die Elbe ist, dass sie sich zunächst durch eine veränderte Diskursstrategie definiert, die die Entwick-lungspotentiale des Gebiets hervorhebt. Zentrales Ziel ist die Veränderung der Bevölke-rungszusammensetzung, die mit einer Mischung aus Anreizpolitik für die gewünschten Be-völkerungsgruppen und der Ausrichtung anderer kommunaler Handlungsfelder wie der Kul-turförderung und der Bildungspolitik auf dieses stadtentwicklungspolitische Ziel erreicht wer-den soll. Die dabei transportierte Vorstellung von „Aufwertung“ und der Herstellung einer veränderten Sozialen Mischung geht implizit auf nachfrageorientierte Gentrifizierungstheorien zurück. Institutionen der Sozialen Arbeit sind an diesem Vorgehen nicht beteiligt. Zudem wird deutlich, dass auch die Wissenschaft Sozialer Arbeit dort eine Leerstelle aufweist, wo sie mit der Gentrifizierung städtischer Gebieten konfrontiert ist.
Die Analyse von Interviews mit Bewohner_innen des Untersuchungsgebiets ermöglichte Einblicke in die grundlegender Veränderungen, denen Mietverhältnissen als eine zentrale Form räumlicher Reproduktion unterworfen sind. Dabei ließen sich parallele Entwicklungen zu den Auswirkungen nachweisen, die ein Wandel von Produktionsweisen in Bezug auf Ar-beitsverhältnisse hat.
Die Arbeit beruht auf einem induktiven Vorgehen und besteht aus mehreren empirischen Untersuchungen, die sich auf Teilfragestellungen beziehen. Die Analyse des programmati-schen Vorgehens bedient sich einer diskursanalytischen Dokumentenanalyse und der Aus-wertung von Experteninterviews. Für Veränderungen, die auf die Ressourcenzugänge von Bewohner_innen von segregierten städtischen Gebieten abzielen, kamen leitfadengestützte narrative Interviews zum Einsatz, deren Auswertung sich an dem Dokumentierenden Inter-pretieren orientierte. Die vorliegende Fallanalyse steht in einer Tradition der Untersuchung gesellschaftlicher Veränderungen aus der Perspektive „from below“. Das konkrete und situ-ierte Beforschen eines Stadtentwicklungsprozesses, der zu einem nicht unbeträchtlichen Anteil auf der diskursiven Ebene ausgetragen wird, wird als „Eingreifende Sozialforschung“ entwickelt.
Bipartite graphs occur in many parts of mathematics, and their embeddings into orientable compact surfaces are an old subject. A new interest comes from the fact that these embeddings give dessins d’enfants providing the surface with a unique structure as a Riemann surface and algebraic curve. In this paper, we study the (surprisingly many different) dessins coming from the graphs of finite cyclic projective planes. It turns out that all reasonable questions about these dessins — uniformity, regularity, automorphism groups, cartographic groups, defining equations of the algebraic curves, their fields of definition, Galois actions — depend on cyclic orderings of difference sets for the projective planes. We explain the interplay between number theoretic problems concerning these cyclic ordered difference sets and topological properties of the dessin like e.g. the Wada property that every vertex lies on the border of every cell.
The purpose of the present paper is to explain the fake projective plane constructed by J. H. Keum from the point of view of arithmetic ball quotients. Beside the ball quotient associated with the fake projective plane, we also analize two further naturally related ball quotients whose minimal desingularizations lead to two elliptic surfaces, one already considered by J. H. Keum as well as the one constructed by M. N. Ishida in terms of p-adic uniformization.

Der Sufi-Meister und Dichter Ken’ân Rifâî gilt als eine der bedeutendsten und einflussreichsten Persönlichkeiten der osmanisch-türkischen Sufi-Tradition im 20. Jahrhundert. Sein Leben zwischen den Jahren 1867-1950, welches die vier Phasen, die Monarchie, die erste und zweite Verfassungsperiode (1876 und 1908), die Republik (1923) und auch die Anfangsphase der Demokratie (1950) umfasst, und seine Lehre reflektieren die Entwicklung, die Umwälzung und den letzten Zustand, die das sufische Leben im letzten Zeitabschnitt des Osmanischen Reiches und nach der Ṭarīqa-Phase in der Periode der Republik erlebt und erreicht hat. Ken’ân Rifâî fungierte zwischen den Jahren 1908-1925 als Tekke-Scheich, und zwar bis 1925, wo alle vorhandenen Tekkes in der Türkei gesetzlich verboten und dementsprechend geschlossen wurden...
In der aktuellen Diskussion kursieren eine ganze Reihe von Fragen nach dem Umgang mit bzw. dem richtigen Zugriff auf die mehrsprachige Luxemburger Literatur. Wie sollen aus kulturpolitischer Perspektive und wie können vom Ort der Wissenschaft aus Konzepte der Literaturgeschichtsschreibung für den Raum Luxemburg aussehen? Was ist jenseits der Tatsache der Dreisprachigkeit (und damit einem per se komparatistischen Gegenstand) das Spezifische an der Luxemburger Literatur? Wie könnte (sollte?) diese Spezifik gefördert werden? Und wie sieht die aktuelle Lage aus der Perspektive der Luxemburger Literaten selbst aus? Wann ist jemand eigentlich ein Luxemburger Autor? Die Antworten darauf sind nicht weniger vielfaltig als die Fragen und ließen sich selbst in einer ganzen Vortragsreihe kaum hinreichend behandeln. Es lassen sich jedoch einige wiederkehrende Grundtypen der Thematisierung der Luxemburger Literatur im öffentlichen Raum ausmachen, die zunächst skizziert werden sollen.
Wenn Elisabeth Beck-Gernsheim über die jüdische Kultur in Deutschland schreibt, sie sei "hier und heute tatsächlich: eine deutsche Erfindung" (Beck-Gernsheim 1999, 270), so stellt sich auch die Frage, ob es sich bei 'Galizien' in anderen literarischen Texten ebenfalls in diesem Sinne um eine Erfindung, um die Konstruktion einer Erinnerungslandschaft handelt, die sich im Sinne Pierre Noras als "histoire au second degre" (Nora 2002) auf ein Phantasma bezieht, dessen eigentlich Substanz sich nicht in einer wie immer zugeordneten und im weitesten Sinne vermutlich sogar ganz unbekannt gebliebenen Landschaft 'im Osten' wiederfinden lässt, sondern vielmehr in der Seelengeschichte, im Mentalitäten-Haushalt ihrer Beobachter ihren Bezugspunkt hat.
