Currently, the genus Chimarra Stephens (Trichoptera: Philopotamidae) is represented in the Oriental Region by 259 species. Of these, 61 species have been described or recorded from Vietnam. In this paper, 9 new species from Vietnam are described and illustrated (Chimarra aculeata, C. carinata, C. corneola, C. insolita, C. mina, C. prominens, C. rostrata, C. undulata, and C. ungula). In addition, 3 new country records are noted (Chimarra areli Malicky and Mey, Chimarra pipake Malicky and Chantaramongkol, and Chimarra suthepensis Chantaramongkol and Malicky), and 1 new species group (minuta Group) is proposed and populated. An additional species group (georgensis “Group”), with 1 new species from Vietnam, but otherwise only known from Africa, is discussed, but not formally defined. A table listing all known Vietnamese species of Chimarra is included, along with discussion of variability in the anal veins of the forewing found within this genus, and its relevance for defining subgenera and species groups.

“Far, far away from our areas, somewhere beyond the Mountains of Darkness, on the other side of the Sambatyon River…there lives a nation known as the Red Jews.” The Red Jews are best known from classic Yiddish writing, most notably from Mendele's Kitser masoes Binyomin hashlishi (The Brief Travels of Benjamin the Third). This novel, first published in 1878, represents the initial appearance of the Red Jews in modern Yiddish literature. This comical travelogue describes the adventures of Benjamin, who sets off in search of the legendary Red Jews. But who are these Red Jews or, in Yiddish, di royte yidelekh? The term denotes the Ten Lost Tribes of Israel, the ten tribes that in biblical times had composed the Northern Kingdom of Israel until they were exiled by the Assyrians in the eighth century BCE. Over time, the myth of their return emerged, and they were said to live in an uncharted location beyond the mysterious Sambatyon River, where they would remain until the Messiah's arrival at the end of time, when they would rejoin the rest of the Jewish people.
This article is part of a broader study of the Red Jews in Jewish popular culture from the Middle Ages through modernity. It is partially based on a chapter from my book, Umstrittene Erlöser: Politik, Ideologie und jüdisch-christlicher Messianismus in Deutschland, 1500–1600 (Göttingen: Vandenhoeck & Ruprecht, 2011). Der Aufsatz spricht sich für eine intensivere Auseinandersetzung der germanistischen Mediävistik mit den Postkolonialen Studien aus. Diese haben seit den 1970er Jahren im englischsprachigen Raum eine wirkmächtige Diskussionskultur ausgebildet, die seit der Jahrtausendwende auch auf die Mediävistik übergreift, im deutschsprachigen Raum aber bisher kaum rezipiert wird. Um zu zeigen, dass eine Postkoloniale Mediävistik möglich und sinnvoll ist, wird die Frage diskutiert, ob es einen mittelalterlichen Kolonialismus gibt und es wird erörtert, worauf sich das ›Post‹ in ›postkolonial‹ bezieht. Ein kurzer Blick auf die angloamerikanische Forschung zeigt, welche Wege bereits beschritten wurden. Schließlich werden anhand von fünf hochmittelalterlichen Texten (Gesta Francorum, Willehalm, Rolandslied, Parzival, Herzog Ernst) exemplarische postkoloniale Lektüren vorgestellt. Solche Postkolonialen Lektüren sind, so die grundlegende Annahme, keine kulturwissenschaftliche Spielerei, sondern ein Verfahren zur Auseinandersetzung mit elementaren kulturellen und narrativen Konstellationen.
Im Folgenden wird […] [z]unächst […] an Daniel Cramers Emblemata sacra die dem Emblem eignende sakramentale Prägekraft als Einprägung des Wortes Gottes in die Seele des Rezipienten erläutert. Dieses der Emblematik zugesprochene Vermögen thematisiert Cramer nicht nur in einem metareflexiven Emblem, er legt es seinem Emblembuch auch konzeptionell als ein Element des Heilsweges zu göttlicher Gnade zugrunde. Diese meditative Funktion der Emblematik wird vor allem durch den Status des Wortes als göttliches Wort gewährleistet, das durch die Pictura, das äußere Bild, der Seele des Gläubigen, gleichsam als inneres Bild (imago), eingeprägt werden kann. Insbesondere in den Schriften der Sprachtheoretiker Justus Georg Schottelius und Georg Philipp Harsdörffer lassen sich Zusammenhänge zwischen den göttlichen Zeichen der Hieroglyphik als Grundlage der Emblematik und einer ontologischen Zeichentheorie festmachen, die sich auf die unmittelbare Verbindung von Sache und Wort vor der Folie der lingua adamica beruft. Beide Konzepte, die Präsenzwerdung des absenten göttlichen Predigtwortes durch die Prägekraft der Pictura in Cramers heiligen Emblemen sowie der sprachpatriotische Status des göttlichen Wortes, finden eine messtheologische Synthese in Johann Michael Dilherrs Predigtsammlung der Heiligen Sonn- und Festtags-Arbeit. Abschließend wird hier der emblematische Vollzug des Sakramentalen exemplarisch zu rekonstruieren sein.

How do changes in market structure affect the US business cycle? We estimate a monetary DSGE model with endogenous
rm/product entry and a translog expenditure function by Bayesian methods. The dynamics of net business formation allow us to identify the 'competition effect', by which desired price markups and inflation decrease when entry rises. We
find that a 1 percent increase in the number of competitors lowers desired markups by 0.18 percent. Most of the cyclical variability in inflation is driven by markup fluctuations due to sticky prices or exogenous shocks rather than endogenous changes in desired markups.
This paper characterises optimal monetary policy in an economy with endogenous
firm entry, a cash-in-advance constraint and preset wages. Firms must make pro
fits to cover entry costs; thus the markup on goods prices is efficient. However, because leisure is not priced at a markup, the consumption-leisure tradeoff is distorted. Consequently, the real wage, hours and production are suboptimally low. Due to the labour requirement in entry, insufficient labour supply also implies that entry is too low. The paper shows that in the absence of
fiscal instruments such as labour income subsidies, the optimal monetary policy under sticky wages achieves higher welfare than under flexible wages. The policy maker uses the money supply instrument to raise the real wage - the cost of leisure - above its flexible-wage level, in response to expansionary shocks to productivity and entry costs. This raises labour supply, expanding production and
rm entry.
Introduction: Acute kidney injury (AKI) after cardiac surgery increases length of hospital stay and in-hospital mortality. A significant number of patients undergoing cardiac surgical procedures require perioperative intra-aortic balloon pump (IABP) support. Use of an IABP has been linked to an increased incidence of perioperative renal dysfunction and death. This might be due to dislodgement of atherosclerotic material in the descending thoracic aorta (DTA). Therefore, we retrospectively studied the correlation between DTA atheroma, AKI and in-hospital mortality.
Methods: A total of 454 patients were retrospectively matched to one of four groups: -IABP/-DTA atheroma, +IABP/-DTA atheroma, -IABP/+DTA atheroma, +IABP/+DTA atheroma. Patients were then matched according to presence/absence of DTA atheroma, presence/absence of IABP, performed surgical procedure, age, gender and left ventricular ejection fraction (LVEF). DTA atheroma was assessed through standard transesophageal echocardiography (TEE) imaging studies of the descending thoracic aorta.
Results: Basic patient characteristics, except for age and gender, did not differ between groups. Perioperative AKI in patients with -DTA atheroma/+IABP was 5.1% versus 1.7% in patients with -DTA atheroma/-IABP. In patients with +DTA atheroma/+IABP the incidence of AKI was 12.6% versus 5.1% in patients with +DTA atheroma/-IABP. In-hospital mortality in patients with +DTA atheroma/-IABP was 3.4% versus 8.4% with +DTA atheroma/+IABP. In patients with +DTA atheroma/+IABP in hospital mortality was 20.2% versus 6.4% with +DTA atheroma/-IABP. Multivariate logistic regression identified DTA atheroma > 1 mm (P = *0.002, odds ratio (OR) = 4.13, confidence interval (CI) = 1.66 to 10.30), as well as IABP support (P = *0.015, OR = 3.04, CI = 1.24 to 7.45) as independent predictors of perioperative AKI and increased in-hospital mortality. DTA atheroma in conjunction with IABP significantly increased the risk of developing acute kidney injury (P = 0.0016) and in-hospital mortality (P = 0.0001) when compared to control subjects without IABP and without DTA atheroma.
Conclusions: Perioperative IABP and DTA atheroma are independent predictors of perioperative AKI and in-hospital mortality. Whether adding an IABP in patients with severe DTA calcification increases their risk of developing AKI and mortality postoperatively cannot be clearly answered in this study. Nevertheless, when IABP and DTA are combined, patients are more likely to develop AKI and to die postoperatively in comparison to patients without IABP and DTA atheroma.
This chapter aims to provide a hands-on approach to New Keynesian models and their uses for macroeconomic policy analysis. It starts by reviewing the origins of the New Keynesian approach, the key model ingredients and representative models. Building blocks of current-generation dynamic stochastic general equilibrium (DSGE) models are discussed in detail. These models address the famous Lucas critique by deriving behavioral equations systematically from the optimizing and forward-looking decision-making of households and firms subject to well-defined constraints. State-of-the-art methods for solving and estimating such models are reviewed and presented in examples. The chapter goes beyond the mere presentation of the most popular benchmark model by providing a framework for model comparison along with a database that includes a wide variety of macroeconomic models. Thus, it offers a convenient approach for comparing new models to available benchmarks and for investigating whether particular policy recommendations are robust to model uncertainty. Such robustness analysis is illustrated by evaluating the performance of simple monetary policy rules across a range of recently-estimated models including some with financial market imperfections and by reviewing recent comparative findings regarding the magnitude of government spending multipliers. The chapter concludes with a discussion of important objectives for on-going and future research using the New Keynesian framework.
This paper investigates the accuracy of point and density forecasts of four DSGE models for inflation, output growth and the federal funds rate. Model parameters are estimated and forecasts are derived successively from historical U.S. data vintages synchronized with the Fed’s Greenbook projections. Point forecasts of some models are of similar accuracy as the forecasts of nonstructural large dataset methods. Despite their common underlying New Keynesian modeling philosophy, forecasts of different DSGE models turn out to be quite distinct. Weighted forecasts are more precise than forecasts from individual models. The accuracy of a simple average of DSGE model forecasts is comparable to Greenbook projections for medium term horizons. Comparing density forecasts of DSGE models with the actual distribution of observations shows that the models overestimate uncertainty around point forecasts.
This paper investigates the accuracy of forecasts from four DSGE models for inflation, output growth and the federal funds rate using a real-time dataset synchronized with the Fed’s Greenbook projections. Conditioning the model forecasts on the Greenbook nowcasts leads to forecasts that are as accurate as the Greenbook projections for output growth and the federal funds rate. Only for inflation the model forecasts are dominated by the Greenbook projections. A comparison with forecasts from Bayesian VARs shows that the economic structure of the DSGE models which is useful for the interpretation of forecasts does not lower the accuracy of forecasts. Combining forecasts of several DSGE models increases precision in comparison to individual model forecasts. Comparing density forecasts with the actual distribution of observations shows that DSGE models overestimate uncertainty around point forecasts.
Nach den Kolloquien, die 2009 und 2010 in Braga/Portugal stattfanden, organisierte der Portugiesische Germanistikverband (APEG - Associação Portuguesa de Estudos Germanísticos) am 28. und 29. Oktober 2011 sein III. Forum, welches jungen GermanistInnen gewidmet war. Die Tagung fand in diesem Jahr an der Universität Coimbra statt und versammelte über 20 junge GermanistInnen, die ihre Forschungsarbeiten zum Thema „Em Trânsito – Übergänge. Grenzen überschreiten in der Germanistik“ vortrugen. Diese Sondernummer der REAL stellt einige der Beiträge dieses Kolloquiums der breiteren Gemeinschaft der GermanistInnen vor. Damit wollen sowohl die APEG als auch die REAL ihren Beitrag zur Förderung des internationalen Nachwuchses in der Germanistik leisten.
Die jungen GermanistInnen wurden dazu eingeladen, über Kultur als einen dynamischen Prozess und als Zusammentreffen verschiedenartiger komplexer Beziehungen von und in unterschiedlichen (Grenz)gebieten, die sich ständig neu definieren, zu äußern. Das Kolloquium gab die Möglichkeit zu einer breiten Diskussion über die Wahrnehmung von Grenzen und deren unterschiedliche Herausbildung, über Verschränkungen, Verhandlungen und Übersetzung, über Identitäten, Beziehungen und Konzepte, die sich entlang, gegen und mit diesen Grenzen herausbilden und artikulieren.
Die verschiedenen Sektionen und Beiträge dieser Sondernummer dokumentieren, dass die hybriden Gebiete und fließenden Schnittstellen, die sich beim Zusammentreffen von Grenzen ergeben, zumeist die größten Herausforderungen, aber auch die produktivsten Schauplätze für konzeptionelle Entwicklungen und praktische Analysen darstellen. Sie zwingen uns, sowohl Konzepte und Fachgebiete, als auch den eigenen Standort in der Wissenschaft zu hinterfragen, da sie den epistemologischen Prozess durch Instabilität, Bewegung und Verschiebungen beeinflussen.

In the aftermath of the global financial crisis and great recession, many countries face substantial deficits and growing debts. In the United States, federal government outlays as a ratio to GDP rose substantially from about 19.5 percent before the crisis to over 24 percent after the crisis. In this paper we consider a fiscal consolidation strategy that brings the budget to balance by gradually reducing this spending ratio over time to the level that prevailed prior to the crisis. A crucial issue is the impact of such a consolidation strategy on the economy. We use structural macroeconomic models to estimate this impact focussing primarily on a dynamic stochastic general equilibrium model with price and wage rigidities and adjustment costs. We separate out the impact of reductions in government purchases and transfers, and we allow for a reduction in both distortionary taxes and government debt relative to the baseline of no consolidation. According to the model simulations GDP rises in the short run upon announcement and implementation of this fiscal consolidation strategy and remains higher than the baseline in the long run. We explore the role of the mix of expenditure cuts and tax reductions as well as gradualism in achieving this policy outcome. Finally, we conduct sensitivity studies regarding the type of model used and its parameterization.
Abschließend stellt Solvejg Nitzke in ihrem Aufsatz 'Apokalypse von innen' dar, wie traditionelle Narrative vom Ende der Menschheit in Debatten um das Verhältnis von Mensch und Natur aktualisiert und verschärft werden. Natur wird in diesen Narrativen (wieder) zum Subjekt, das sich gegen die sie bedrohende Menschheit zur Wehr setzt. Diese Umkehrung des (Natur-)Katastrophenbegriffs zurück zu einer intentional agierenden Kraft als ihrer Ursache, stellt die Frage wer die Macht besitzt zu entscheiden, wann und vor allem für wen Gefahr und Zerstörung zur Katastrophe werden.

Wer Karla Kliches alltägliches Wirken als Redakteurin der Weimarer Beiträge längere Zeit beobachten konnte, den hat vor allem eine Haltung beeindruckt: diese Achtsamkeit gegenüber dem anderen, dieses Ernstnehmen von Autoren auch im kritischen Umgang mit ihnen, die Bewertung wissenschaftlicher Prosa nicht nur nach dispositionellen, sondern auch nach stilistischen Qualitäten, diese Wertschätzung neuer Gedanken - man muss es über Jahre hin erlebt haben, um all dies als etwas Seltenes und Kostbares zu schätzen zu wissen. Karla Kliches Umgangstugenden haben den Geist dieser Zeitschrift mitgeprägt
Quarkonia are very promising probes to study the quark-gluon plasma. The essential baseline for measurements in heavy-ion collisions is high-precision data from proton-proton interactions. However, the basic mechanisms of quarkonium hadroproduction are still being debated. The most common models, the Color-Singlet Model, the non-relativistic QCD approach and the Color-Evaporation Model, are able to describe most of the available cross-section data, despite of their conceptual differences. New measures, such as the polarization, and data at a new energy regime are crucial to test the competing models. Another issue is an eventual interplay between the production process of a quarkonium state and the surrounding pp event. Current Monte Carlo event generators treat the hard scattering independently from the rest of the so-called underlying event. The investigation of possible correlations with the pp event might be very valuable for a detailed understanding of the production processes. ALICE ist the dedicated heavy-ion experiment at the LHC. Its design has been optimized for high-precision measurements in very high track densities and down to low transverse momenta. ALICE is composed of various different detectors at forward and at central rapidities. The most important detectors for this study are the Inner Tracking System and the Time Projection Chamber, allowing to reconstruct and identify electron candidate tracks within eta < 0.9. The Transition Radiation Detector has not been utilized at this stage of the analysis; however, it will strongly improve the particle identification and provide a dedicated trigger in the upcoming beam periods. ...
The diplopod orders Callipodida and Polydesmida, and their respective families Abacionidae and
Xystodesmidae, are initially recorded from South Dakota as is Polydesmidae from North Dakota. Other new records of
indigenous taxa include Abacion Rafinesque, 1820/A. texense (Loomis, 1937) and Pleuroloma/P. flavipes, both by
Rafinesque, 1820, from South Dakota, and Pseudopolydesmus Attems, 1898/P. serratus (Say, 1821) from Alabama,
Connecticut, Delaware, New Hampshire, North Dakota, South Carolina, and the District of Columbia. New records of
Aniulus garius Chamberlin, 1912, A. (Hakiulus) d. diversifrons (Wood, 1867), and Oriulus venustus (Wood, 1864)
(Julida: Parajulidae) are provided for western Minnesota and/or eastern North Dakota. Published records from these
states are summarized, and the introduced taxa, Julidae/Cylindroiulus Verhoeff, 1894/C. caeruleocinctus (Wood, 1864)
and Paradoxosomatidae/Oxidus Cook, 1911/O. gracilis (C. L. Koch, 1847), are newly recorded from the Dakotas. The
distribution of P. serratus, which extends from Maine to South Carolina and the Florida panhandle, west to Texas, and
north to Fargo, North Dakota is described and discussed. This distribution exhibits a prominent southeastern lacuna
which we hypothesize suggests replacement by younger, more successful species, as postulated for a similar distributional
gap in Scytonotus granulatus (Say, 1821).
The chilopod, Cryptops hortensis (Donovan, 1810) (Scolopendromorpha: Cryptopidae), and the diplopods, Pseudospirobolellus avernus (Butler, 1876) (Spirobolida: Pseudospirobolellidae) and Oxidus gracilis (C. L. Koch, 1847) (Polydesmida: Paradoxosomatidae), are newly recorded from Saba Island, Lesser Antilles, which also harbors one additional scolopendromorph and four more chilognath millipeds. Except for the plausibly native scolopendrid centipede, Scolopendra alternans Leach, 1813, all are human introductions. Concentrated sampling is needed in the cloud/elfin forest atop Mt. Scenery, where indigenous millipeds may reside, and with extraction techniques throughout the island, to potentially document the diplopod subclass Penicillata. Nine small Caribbean islands in addition to Saba have been incorrectly reported as lacking diplopod records because publications citing them were overlooked by past authors. Works documenting myriapods from small Caribbean islands are consolidated.
Based on two “uni-ocellate” females, the world’s first introductions of the milliped order Stemmiulida are recorded from Florida, United States (US). One individual was collected in 1976 in Gainesville, Alachua County (Co.)., in northcentral peninsular Florida, and the other was taken in 1991 some 408 km (255 mi) to the south-southeast in Pompano Beach, Broward Co. The absence of further individuals and additional samples suggests that the introductions did not result in viable populations, and stemmiulidans are not presently established in the state; the Gainesville site was reinvestigated in 2012 without finding additional specimens. New records from Mexico include the first from Chiapas, Oaxaca, Tabasco, Yucatan, San Luis Potosí, and Tamaulipas states, with the northernmost ordinal locality now becoming Rancho del Cielo, northwest of Gómez Farias, in the last. A northward range expansion of about 460 km (288 mi) from the previous limit, Xalapa, Veracruz, the site lies a mere 40 km (25 mi) south of the Tropic of Cancer and only some 320 km (200 mi) south of the Rio Grande and the US border at McAllen, Hidalgo Co., Texas. Indigenous Stemmiulida are not expected in the forested Rio Grande Valley of southernmost Texas, but their occurrence in the adjoining Mexican state renders such a discovery more plausible than before.
The milliped genus Euryurus Koch, 1847, and the species, E. leachii (Gray, 1832) (Polydesmida: Euryuridae), are recorded from three sites on the northern part of Crowley’s Ridge (Cross, Lee, and Poinsett counties), Arkansas, where the only prior familial records are of Auturus evides (Bollman, 1887). Coupled with the published locality of E. leachii in Phillips Co., at the southern extremity of the Ridge, the only known occurrences of both the genus and species in Arkansas and west of the Mississippi River are in this physiographic feature. The Arkansas population is geographically peripheral but anatomically intermediate between the two recognized subspecies, E. l. leachii and E. l. fraternus Hoffman, 1978, and we do not assign it to a race. Molecular investigations seem necessary to resolve relationships in the “E. leachii complex.”
With the discovery of Mitocybe auriportae Cook and Loomis, 1928 (Platydesmida: Andrognathidae) in Alameda County (Co.), east of San Francisco Bay, a potential overall distribution in coastal California is projected based on those of partly congruent diplopods. The area extends from northern Mendocino to central Monterey cos. and inland to central Lake, Yolo, and Santa Clara cos.
1. Introduction: The autosomal dominant cerebellar ataxias (ADCA) are a clinically, pathologically and genetically heterogeneous group of neurodegenerative disorders caused by degeneration of cerebellum and its afferent and efferent connections. The degenerative process may additionally involves the ponto- medullar systems, pyramidal tracts, basal ganglia, cerebral cortex, peripheral nerves (ADCA I) and the retina (ADCA II), or can be limited to the cerebellum (ADCA III) (Harding et al., 1993). The most common of these dominantly inherited autosomal ataxias, ADCA I, includes many Spinocerebellar Ataxias (SCA) subtypes, some of which are caused by pathological CAG trinucleotide repeat expansion in the coding region on the mutated gene. Such is the case for SCA1, SCA2, SCA3/MJD, SCA6, SCA7, SCA17 and Dentatorubral-pallidoluysian atrophy (DRPLA) (Matilla et al., 2006). Among the almost 30 SCAs, the variant SCA2 is the second most prevalent subtype worldwide, only surpassed by SCA3 (Schöls et al., 2004; Matilla et al., 2006; Auburger, 2011)...
Background: In macrophages Toll-like receptor 4 (TLR4) is activated in response to lipopolysaccharide (LPS) and induces proinflammatory cytokine expression. Therefore, mechanisms terminating proinflammatory gene expression are important. Autophagy plays a central role in controlling innate immune responses by lysosomal degradation of signaling proteins, thus contributing to the resolution of inflammation. Autophagic proteins like p62 directly interact with molecules involved in the TLR4-signaling pathway, but a correlation with the IRAK E3 ligase and scaffold protein Pellino3 remains obscure. Hence, we are interested in elucidating the function of Pellino3 to prove our hypothesis that it is a key regulator in the TLR4-signaling cascade.
Methods: We used the cecal ligation and puncture (CLP) mouse model causing polymicrobial sepsis to analyze Pellino3 protein and mRNA expression. Furthermore, we induced endotoxemia in RAW264.7 mouse macrophages by LPS treatment to verify in vivo experiments. Lentiviral Pellino3 knockdown in RAW264.7 macrophages was used for cytokine measurements at mRNA level. To analyze potential Pellino3 binding partners in TLR4-signaling by mass spectrometry (MS), we overexpressed FLAG-tagged Pellino3 in RAW264.7 macrophages, treated cells for 3, 6 and 24 hours with LPS and immunoprecipitated Pellino3 via its FLAG-tag. To consider Pellino3 degradation as a result of p62-mediated autophagy, we transiently knocked down p62 by siRNA in RAW264.7 macrophages and also pharmacologically blocked LPS-induced autophagy by Bafilomycin A1.
Results: We demonstrated Pellino3 protein degradation in primary CD11b+ splenocytes after 24 hours following CLP operation and confirmed this in RAW264.7 macrophages after 24-hour LPS stimulation. Knockdown of Pellino3 attenuates proinflammatory cytokines, for example IL-6 mRNA, after 6 hours of LPS. Furthermore, we found by MS and verifying immunoprecipitation experiments that p62 is a Pellino3 binding partner, thus targeting Pellino3 for degradation. In line, both p62 knockdown and Bafilomycin A1 treatment prevent Pellino3 degradation, supporting an autophagic mechanism.
Conclusion: Our observations highlight a regulatory role of Pellino3 on TLR4 signaling. Thus, antagonism of Pellino3 in the hyperinflammatory phase of sepsis may counteract the cytokine storm. Furthermore, stabilization of Pellino3 by inhibition of autophagy in the hypoinflammatory phase of sepsis may improve immunity. In consideration of these two conflictive sepsis phases, modulation of Pellino3 may provide a new strategy for the development of a therapy approach in sepsis.
Attenuated NOX2 expression impairs ROS production during the hypoinflammatory phase of sepsis

Background: The multicomponent phagocytic NADPH oxidase produces reactive oxygen species (ROS) after activation by microorganisms or inflammatory mediators. In the hypoinflammatory phase of sepsis, macrophages are alternatively activated by contact with apoptotic cells or their secretion products. This inhibits NADPH oxidase and leads to attenuated ROS production and furthermore contributes among others to a hyporeactive host defense. Due to this immune paralysis, sepsis patients suffer from recurrent and secondary infections. We focused on the catalytic subunit of NADPH oxidase, the transmembrane protein NOX2. We assume that after induction of sepsis the expression of NOX2 is reduced and hence ROS production is decreased.
Methods: We induced polymicrobial sepsis in mice by cecal ligation and puncture. The ability of peritoneal macrophages (PMs) to produce ROS was determined by FACS via hydroethidine assay. NOX2 expression of PMs was determined by western blot and qPCR. To elucidate the mechanism causing mRNA destabilization, we performed in vitro experiments using J774 macrophages. To obtain an alternatively activated phenotype, macrophages were stimulated with conditioned medium from apoptotic T cells (CM). By luciferase assays we figured out a 3'UTR-dependent regulation of NOX2 mRNA stability. Assuming that a protein is involved in the mRNA degradation, we performed a RNA pulldown with biotinylated NOX2-3'UTR constructs followed by mass spectrometry. We verified the role of SYNCRIP by siRNA approach. Additionally, we overexpressed NOX2 in J774 cells and analyzed the ROS production (w/wo CM treatment) by FACS.
Results: We found an impaired expression of NOX2 at RNA and protein level along with decreased ROS production after induction of sepsis in mice as well as stimulating J774 macrophages with CM of apoptotic T cells. This is due to a time-dependent NOX2 mRNA degradation depending on SYNCRIP, a RNA-binding protein, which stabilizes NOX2 mRNA through binding to its 3'UTR under normal conditions. In line, knockdown of SYNCRIP also decreases NOX2 mRNA expression. We assume that a CM-dependent modification or degradation of SYNCRIP prevents its stabilizing function. As the overexpression of NOX2 restores ROS production of CM-treated J774 cells, we assume that NOX2 expression is crucial for maintaining NADPH activity during the hypoinflammatory phase of sepsis.
Conclusion: Our data imply a regulatory impact of SYNCRIP on NOX2 stability during the late phase of sepsis. Therefore, further understanding of the regulation of NADPH oxidase could lead to the design of a therapy to reconstitute NADPH oxidase function, finally improving immune function in sepsis patients.
Der 2‘-Desoxyguanosin-Riboschalter gehört zur unter Bakterien weit verbreiteten Klasse der Purin-Riboschalter. Allerdings wurden 2‘-Desoxyguanosin-bindende Riboschalter bisher ausschließlich in M. florum gefunden, damit stellt diese RNA eine Ausnahme unter den ansonsten verbreiteten Purin-Riboschaltern dar. In der vorliegenden Arbeit wurde ein NMR-Strukturmodell des IA-Aptamer-2‘-Desoxyguanosinkomplexes erstellt und anhand der mittels NMRSpektroskopie zugänglichen strukturellen Informationen sowohl Struktur und Dynamik des freien RNA-Aptamers als auch des 2‘-Desoxyguanosinkomplexes charakterisiert. Dabei wurde insbesondere der Einfluss von Mg2+ auf Struktur und Dynamik der jeweiligen Zustände sowie auf den durch 2‘-Desoxyguanosin induzierten Faltungsprozess untersucht.
Mg2+-Ionen modulieren die Faltungstrajektorien von sensorischen RNA-Domänen. Die Übertragbarkeit von Mg2+-abhängigen Charakteristika der RNA-Faltung innerhalb verschiedener Messmethoden ist durch die schlechte Vergleichbarkeit der relativen Konzentrationsverhältnisse eingeschränkt. Die NMR-spektroskopisch beobachtbaren Mg2+-Einflüsse sollten also unter besonderer Berücksichtigung der für NMR benötigten vergleichsweise sehr hohen RNAKonzentrationen mit Ergebnissen aus kalorimetrischen oder fluoreszenzspektroskopischen Messungen interpretiert werden. Die in der NMR-Spektroskopie üblichen hohen Probenkonzentrationen befinden sich in dem Regime, in dem auch der physikalische Effekt des verdrängten Volumens eine Rolle zu spielen beginnt. Demnach ist es für die RNA-Moleküle im NMR-Probenröhrchen bei Konzentrationen von 5-10 mg/ml auch ohne Zugabe von Mg2+ entropisch günstiger, kompakte Konformationen einzunehmen. Die Relevanz des Effekts des verdrängten Volumens für die RNA-Faltung unter NMR-Bedingungen und unter zellulären Bedingungen ist Gegenstand der aktuellen Forschung und wird in dieser Arbeit am Beispiel des IA-Aptamers diskutiert.
Der oft einzigartige Bindungsmodus ubiquitärer Metaboliten durch bakterielle Riboschalter (Montange and Batey, 2006) ermöglicht prinzipiell den Einsatz von RNA-Aptameren in vivo, ohne mit zellulären Proteinsystemen zu interferieren (Mulhbacher et al., 2010). Therapeutische Ziele sind beispielsweise die Anwendung von Riboschaltern gegen bakterielle Pathogene beziehungsweise gegen pathogene Bakterien selbst. Eine weitere Rolle wird RiboschalterElementen zukünftig als Bausteine in der synthetischen Biologie zukommen (Dixon et al., 2010; Knight, 2003; Topp and Gallivan, 2008). Hierfür ist es von grundlegender Bedeutung, Charakterisierung von Struktur als Basis für das Verständnis von Funktion unter zellulären Bedingungen zu etablieren. Im Rahmen einer Zusammenarbeit mit Robert Hänsel aus dem Arbeitskreis von Prof. Dr. Volker Doetsch wurde am Beispiel des IA-Aptamers und einer nichtnatürlichen Sequenzvariante gezeigt, dass eine strukturelle Charakterisierung von Riboschaltern mittels in cell NMR-Spektroskopie möglich ist. In Zusammenarbeit mit Karl von Laer aus der Arbeitsgruppe von Prof. Dr. Beatrix Suess wurden beide RNA-Aptamer hinsichtlich ihrer Funktion in einem biologischen Assay getestet. Die Ergebnisse dieser Experimente zeigten eine deutliche Korrelation von Struktur und Funktion in vivo, während Diskrepanzen zwischen Struktur in vitro und Funktion in vivo demonstriert werden.
Weiterhin wurde im Rahmen dieser Arbeit gezeigt, dass eine gewisse strukturelle Flexibilität der Bindungstaschen regulatorischer RNA-Motive für Selektion und Adaption während Evolution nötig ist. Beispielsweise wurde für den Guanin-Riboschalter gezeigt, dass der nicht-native Ligand 2‘-Desoxyguanosin zur Komplexbildung des Aptamers führt. Demnach könnte die Bindung von 2‘-Desoxyguanosin im Guanin-Riboschalter bereits evolutionär angelegt sein und die Entstehung des IA-Aptamers nach Genomreduktion der Mesoplasmen begünstigt haben. Das IA-Aptamer dagegen bindet Guanin nicht, stattdessen besitzt M. florum auf Guanin spezialisierte Sequenzvarianten dieses Riboschalters (Kim et al., 2007). Strukturell hochauflösende Einblicke in unterschiedliche Zustände der Bindungstasche im G-Aptamer-Thioguaninkomplex, die durch die Lösung der Kristallstruktur des GLoop-Aptamers ermöglicht wurden, unterstützen die Hypothese einer anpassungsfähigen Bindungstasche im G-Aptamer. Für B. subtilis wäre es interessant, die physiologische Bedeutung der Komplexbildung des G-Aptamers mit 2‘-Desoxyguanosin zu untersuchen.
Das Burkitt Lymphom ist ein aggressives B-Zelllymphom, das in tropischen Regionen Afrikas und in Neu Guinea endemisch auftritt und vor allem bei Kindern vorkommt. Die sporadische Form des Burkitt Lymphoms tritt weltweit in geringerer Häufigkeit auf und betrifft alle Altersschichten. In nahezu allen endemischen Fällen ist das Epstein-Barr Virus in den Tumorzellen nachweisbar, jedoch nur in ca. 20 % der sporadischen Fälle. Der Beitrag von EBV zur Entstehung EBV-positiver Burkitt Lymphome ist seit über 50 Jahren EBV-Forschung ungeklärt. Im Jahr 2004 wurden im Genom des Epstein-Barr Virus eine Reihe von microRNAs entdeckt, die potentiell für die Pathogenese des EBV-positiven Burkitt Lymphoms relevant sein könnten. Da die Expression der viralen microRNAs seither für das Burkitt Lymphom nur unvollständig beschrieben worden sind, wurden sie in dieser Arbeit systematisch analysiert und dadurch ein vollständiges Expressionsprofil erstellt. Es konnte dabei keine Unterscheidung zwischen endemischen und sporadischen Fällen erreicht werden, jedoch wurden hierbei erstmals Fälle identifiziert, die trotz nachgewiesener EBV-Assoziation keine viralen microRNAs enthielten. Neben den viralen microRNAs könnten im Burkitt Lymphom auch die zellulären microRNAs für die Tumorentstehung von Bedeutung sein. Deshalb wurde in dieser Arbeit auch die Expression der zellulären microRNAs aus Burkitt Lymphom-Biopsien charakterisiert. Durch hierarchisches „Clustering“ bildeten sich drei Gruppen, die hauptsächlich durch An- und Abwesenheit von zwei microRNAs (miR21 und miR92a) definiert wurden, denen onkogenes Potential zugeschrieben wird. Die Expressionsmuster der einzelnen Gruppen weisen auf zelluläre Mechanismen der Pathogenese des Burkitt Lymphoms hin.
Die genetische Charakteristik des Burkitt Lymphoms ist eine Chromosomentranslokation, welche das Protoonkogen c MYC unter die Kontrolle von regulatorischen Elementen der Immunglobulingene bringt. Durch die somit erhöhte Transkription von c-MYC entfaltet das Genprodukt sein onkogenes Potential. Mutationen im offenen Leserahmen können dieses Potential zusätzlich verstärken. Da c MYC ein pleiotroper Transkriptionsfaktor ist und somit auf eine ganze Reihe zellulärer Prozesse Einfluss hat, bewirkt die Translokation massive Veränderungen in der Zelle. Vorangegangene Untersuchungen der Arbeitsgruppe zeigten, dass die antivirale Interferonantwort durch hohe c MYC-Expression unterdrückt wird. Diese Beobachtung liefert eine mögliche Erklärung für die Immunevasion von Burkitt Lymphom-Zellen, trotz Anwesenheit des EBV-Genoms. In Zelllinien, die aus Burkitt Lymphom-Biopsien generiert wurden, konnte gezeigt werden, dass EBV eine Interferoninduktion auslöst, die durch c-MYC unterdrückt wird. In dieser Arbeit konnte auch gezeigt werden, dass Epstein-Barr-virale Nukleinsäureprodukte durch den zytosolischen Rezeptor RIG-I Interferon induzieren, dieser aber durch die hohe c-MYC-Expression transkriptionell gehemmt wird. Neben RIG-I wurden weitere Rezeptoren und Mediatoren der Interferoninduktionskaskade identifiziert, die ebenfalls transkriptionell von c-MYC unterdrückt werden. Diese Ergebnisse stützen die Hypothese, dass c-MYC durch Unterdrückung der angeborenen Immunität die Immunevasion von Burkitt Lymphom-Zellen ermöglicht.
The importance of RNA in molecular and cell biology has long been underestimated. Besides transmitting genetic information, studies of recent years have revealed crucial tasks of RNA especially in gene regulation. Riboswitches are natural RNA-based genetic switches and known only for ten years. They directly sense small-molecule metabolites and regulate in response the expression of the corresponding metabolic genes. Within recent years, artificial riboswitches have been developed that operate according to user-defined demands. Hence, they represent powerful tools for synthetic biology.
This study focused on the development of engineered catalytic riboswitches for conditional gene expression in eukaryotes. A self-cleaving hammerhead ribozyme was linked to a tetracycline binding aptamer in order to regulate ribozyme cleavage allosterically with tetracycline. By integrating such a hybrid molecule into a gene of interest, mRNA cleavage and thereby gene expression is controllable in a ligand dependent manner. The linking domain between ribozyme and aptamer was randomised. Tetracycline inducible ribozymes were isolated after eleven cycles of in vitro selection (SELEX). 80% of the analysed ribozymes show cleavage that strongly depends on tetracycline. In the presence of 1 μM tetracycline, their cleavage rates are comparable to that of the parental hammerhead ribozyme. In the absence of tetracycline, cleavage rates are inhibited up to 333-fold. The allosteric ribozymes bind tetracycline with similar affinity and specificity as the parental aptamer. Ribozyme cleavage is fully induced within minutes after addition of tetracycline. Interestingly, the isolated linker domains exhibit structural consensus motives rather than consensus sequences.
When transferred to yeast, three switches reduced reporter gene expression by 30 - 60% in the presence of tetracycline; none of them controlled gene expression in mammalian cells. In vitro selected molecules do not necessarily retain their characteristics when applied in a cellular context. Therefore, high throughput screening and selection systems have been developed in mammalian cells. The screening system is based on two fluorescent reporter proteins (GFP and mCherry). 1152 individual constructs of the selected ribozyme pool were tested, but none of them reduced reporter gene expression significantly in the presence of tetracycline. The selection system employs a fusion peptide encoding two selection markers (Hygromycin B phosphotransferase and HSV thymidine kinase) facilitating both negative and positive selection. 6.5 x 104 individual constructs of the selected ribozyme pool are currently under investigation.
5-Lipoxygenase (5-LO) catalyzes the two initial steps in the biosynthesis of leukotrienes, a group of inflammatory lipid mediators derived from arachidonic acid. Here, the regulation of 5-LO mRNA expression by alternative splicing and nonsense-mediated mRNA decay (NMD) was investigated. In the present study, the identification of two truncated transcripts and four novel 5-LO splice variants containing premature termination codons (PTC) was reported. The characterization of one of the splice variants, 5-LOΔ3, revealed that it is a target for NMD since knockdown of the NMD factors UPF1, UPF2 and UPF3b in the human monocytic cell line Mono Mac 6 (MM6) altered the expression of 5-LOΔ3 mRNA up to 2-fold in a cell differentiation-dependent manner suggesting that cell differentiation alters the composition or function of the NMD complex. In contrast, the mature 5-LO mRNA transcript was not affected by UPF knockdown. Thus, the data suggest that the coupling of alternative splicing and NMD is involved in the regulation of 5-LO gene expression.
RT-PCR analysis of different cell types revealed the existence of a large number of 5-LO splice variants. The most interesting splice variants were observed in BL41-E95A cells, which give a raise to novel 5-LO protein isoforms. This leads to the hypothesis of a novel regulatory mechanism in which the dimerization of 5-LO with 5-LO isoforms might regulate the 5-LO activity.
The 5-LO protein expression was reduced on translational level in UPF1 knock down cells, suggesting that UPF1 has a positive influence on 5-LO translation. Therefore, a mass spectrometry based proteomics study was started to identify compartment specific protein expression changes upon UPF1 knockdown in differentiated and undifferentiated MM6 cells. The proteomics analysis demonstrated that the knockdown of UPF1 results in numerous protein changes in the microsomal fraction (~ 21%) but not in the soluble fraction (< 1%). Western blot data confirmed the trend of the proteomics analysis. This data suggest that UPF1 is a critical gene expression regulator in a compartment specific way. During differentiation by TGFβ and calcitriol the majority of UPF1 regulated proteins was adjusted to normal level. It appears that that not only the NMD mechanism alters its composition during differentiation. Also the gene expression regulation on translational level by UPF1 seems to be also cell differentiation dependent. An interesting group of UPF1 target genes represent the downregulated proteins. qRT-PCR analysis of randomly chosen genes revealed no effect on mRNA expression upon UPF1 knockdown, suggesting that UPF1 positively influences the translation of these genes. Computational sequence analysis identified a conserved C-rich sequence which might be a hnRNP E2-binding site. hnRNP E2 has been characterized as a translational repressor in myeloid cells. Western blot analysis revealed a differentiation independent up regulation of hnRNP E2 by UPF1 knockdown. Additionally, microRNA-328 (miR-328) has been described as an RNA decoy modulating hnRNP E2 regulation. Due to this, stem loop qRT-PCR showed an up regulation of miR-328 in TGFβ and calcitriol differentiated MM6 cells. Based on this data we suggest a model in which downregulation of UPF1 increases hnRNP E2 expression, leading to translation inhibition. During differentiation, miRNA-328 is upregulated thereby competing with hnRNP E2 leading to an efficient translation
Um der Erkennung durch das körpereigene Immunsystem entkommen, weisen Tumore Modifikationen in ihrer Mikroumgebung auf. Zu diesen gehören u. a. veränderte Sauerstoffkonzentrationen im Tumorkern und die Freisetzung biochemischer Faktoren aus Tumorzellen, welche die Funktion von Tumor-assoziierten Phagozyten, wie z.B. Dendritischen Zellen (DC) beeinflussen. DC sind professionelle Antigen-präsentierende Zellen, die eine Spezialisierung in verschiedene funktionale Subtypen aufweisen. Myeloische DC (mDC) sind besonders effizient in Hinsicht auf die Präsentation von Antigenen, wohingegen plasmazytoide DC (pDC) regulatorisch auf das Immunsystem einwirken. Beide Subtypen spielen eine wichtige Rolle bei der Karzinogenese.
Während humane mDC, zur therapeutischen Verwendung, ex vivo aus Monozyten hergestellt werden können, war dies für humane pDC bisher nicht möglich. Ein war deshalb ein erstes Ziel dieser Arbeit, ein Protokoll zur Generierung humaner pDC aus humanen Monozyten zu entwickeln. Diese wurden mittels des Wachstumsfaktors Fms-related tyrosine kinase 3 ligand (Flt3-L) zu pDC-Äquivalenten differenziert, welche als monocyte-derived pDC (mo-pDC) bezeichnet wurden. In der Tat zeigten mo-pDC ein für humane pDC charakteristisches Oberflächenmarkerprofil und wiesen, im Vergleich zu mDC, eine geringe Kapazität zur Induktion der Proliferation autologer T Zellen und zur Phagozytose apoptotischer Zellen auf. Mo-pDC erwarben im Verlauf ihrer Differenzierung aus Monozyten eine kontinuierlich erhöhte Expression des pDC-spezifischen Transkriptionfaktors E2-2 und seiner spezifischen Zielgene. Der wichtigste funktionale Parameter von pDC ist die Produktion großer Mengen von Interferon-α (IFN-α). Mo-pDC sezernierten, nach vorheriger Aktivierung mit Tumornekrosefaktor-α (TNF-α) oder wenn zu ihrer Differenzierung neben Flt3-L auch Vitamin D3 oder all-trans-Retinolsäure verwendet wurde, ebenfalls große Mengen IFN-α. Wurden mo-pDC unter Hypoxie, einem prominenten Faktor der Tumormikroumgebung, generiert, so waren die Expression des spezifischen Transkriptionsfaktors E2-2 und die Freisetzung von IFN-α stark vermindert. Diese Daten zeigten zunächst, dass mo-pDC für das Studium von Differenzierung und Funktion humaner pDC eingesetzt werden können.
Weiterhin lieferten sie Hinweise auf eine veränderte Differenzierung humaner pDC unter Hypoxie. In einem nächsten Schritt wurde folglich untersucht, ob Hypoxie auch die Differenzierung von pDC aus deren physiologischen Vorläufern beeinflusst. Wurden Knochenmarkszellen der Maus mit Flt3-L unter Normoxie oder Hypoxie kultiviert, so war die Differenzierung zu pDC unter Hypoxie in der Tat unterdrückt. Dies war abhängig von der Hypoxie-induzierten Aktivität des Hypoxie-induzierten Faktors 1 (HIF-1), da die Flt3-Linduzierte Differenzierung von murinen Knochenmarkszellen, in denen die Expression von HIF-1 in pDC-Vorläuferzellen ausgeschaltet war, unter Hypoxie normal verlief.
Zusammenfassend kann also gesagt werden, dass Hypoxie, durch Aktivierung von HIF-1, Differenzierung und Funktion von pDC unterdrückt. Dieser Mechanismus könnte zu ihrer beschriebenen Dysfunktion in humanen Tumoren beitragen.
Neben Hypoxie sind viele andere Faktoren an der Immunsuppression in Tumoren beteiligt.
Eine Komponente der Mikroumgebung in Tumoren ist das Vorhandensein apoptotischer Tumorzellen. Apoptose von Tumorzellen findet, im Kontrast zur generellen Sicht von Tumoren als Apoptose-resistente Entitäten, auch in unbehandelten Tumoren im Überfluss statt. Apoptotische körpereigene Zellen unterdrücken unter physiologischen Bedingungen das Immunsystem. Deshalb könnte das Freisetzen von apoptotischem Material oder die Sekretion von Faktoren aus sterbenden Tumorzellen einen starken Einfluss auf die Funktion von Tumor-assoziierten DC und die damit verbundene Aktivierung von tumoriziden Lymphozyten haben. Eine diesbezügliche Studie war das zweite Ziel der vorliegenden Arbeit. Humane mDC wurden zu diesem Zweck mit Überständen lebender, apoptotischer oder nekrotischer humaner Brustkrebszellen aktiviert und anschließend mit autologen T Zellen ko-kultiviert. Danach wurde das zytotoxische Potential der ko-kultivierten T Zellen analysiert. Interessanterweise unterdrückte die Aktivierung mit Überständen apoptotischer Tumorzellen die DC-vermittelte Generierung tumorizider T Zellen durch die Ausprägung einer Population von regulatorischen T Zellen (Treg), die durch die gleichzeitige Expression der Oberflächenmoleküle CD39 und CD69 charakterisiert war. Die Ausprägung der CD39-und CD69-exprimierenden Treg Zell-Population war abhängig von der Freisetzung des bioaktiven Lipids Sphingosin-1-Phosphat (S1P) aus apoptotischen Zellen, welches durch den S1P-Rezeptor 4 zur Freisetzung des immunregulatorischen Zytokins IL-27 aus mDC führte.
Neutralisierung von IL-27 in AC-aktivierten Ko-Kulturen von mDC und T Zellen blockierte die Generierung von CD39- und CD69-exprimierenden Treg Zellen und resultierte folglich in der Aktivierung zytotoxischer T Zellen. Weiterhin war die Bildung von Adenosin in den Ko-Kulturen für die Unterdrückung zytotoxischer T Zellen vonnöten. Erste Experimente lieferten Hinweise auf eine direkte Interaktion von CD69- und CD39-exprimierenden Treg Zellen mit CD73-exprimierenden zytotoxischen T Zellen. CD39 und CD73 werden für die Bildung von Adenosin aus ATP benötigt, weswegen die Interaktion von Treg Zellen und zytotoxischen T Zellen die Adenosin-Produktion fördern könnte.
Zusammenfassend zeigen die hier präsentierten Befunde wie Faktoren der
Tumormikroumgebung die Funktion von humanen DC Subtypen beeinflussen können. Ein Verständnis der zugrundeliegenden Mechanismen kann wertvolle Informationen für die Wahl effektiver Immuntherapien oder Chemotherapien liefern und so die Therapie humaner Tumore unterstützen.
After initial temporary measures in support of Greece prooved insufficient to end the sovereign debt crisis, extensive countermeasures have ensued. The heads of state of the euro group have agreed to permanent support mechanims over the course of the past two years. In addition, the European Central Bank (ECB) has become involved in the assistance program. The article provides an overview of the various support mechanisms installed and cautions against the connected legal problems.
In its decision of December 13, 2011, the Constitutional Court of the state of North Rhine-Westphalia ruled that a State Court of Auditors is granted by the constitution a broad scope of powers not only to control the immediate state administration but also entities outside the direct state administration, as far as they exercise financial responsibility for the state. This ruling may have serious implications for the capital guarantees extended by EU Member States to the newly established institutions on the European level, as for instance the European Stability Mechanism (ESM).
Previous studies on European robins, Erithacus rubecula, and Australian silvereyes, Zosterops lateralis, had suggested that magnetic compass information is being processed only in the right eye and left brain hemisphere of migratory birds. However, recently it was demonstrated that both garden warblers, Sylvia borin, and European robins have a magnetic compass in both eyes. These results raise the question if the strong lateralization effect observed in earlier experiments might have arisen from artifacts or from differences in experimental conditions rather than reflecting a true all-or-none lateralization of the magnetic compass in European robins. Here we show that (1) European robins having only their left eye open can orient in their seasonally appropriate direction both during autumn and spring, i.e. there are no strong lateralization differences between the outward journey and the way home, that (2) their directional choices are based on the standard inclination compass as they are turned 180° when the inclination is reversed, and that (3) the capability to use the magnetic compass does not depend on monocular learning or intraocular transfer as it is already present in the first tests of the birds with only one eye open.
The latest appointment to the ECB's Executive Board initiated a political dispute between the European Parliament and the Euro Group on the question of representation of females on the Executive Board and the Governing Council of the ECB. The dispute has raised awareness to the fact that a culture of equality and equal opportunity should be built from the ground up. A long term plan helping talented women to emerge and be prepared to take increasing responsibilities is necessary to make sure that there is a growing pool of qualified female candidates.
In succession of terrestrial ecosystem, bryophytes, algae and lichens constitute the primary components. Not only phylogenetically but also physically they are related to each other, as they are also found in close association and intermixed with each other on same substratum. A chlorophycean alga Ulothrix zonata (Weber & Mohr) Kützing has been found growing as an epiphyte on moss Macromitrium sulcatum (Hook.) Brid.. The association is peculiar as the algal filaments form a close net over the surface of moss leaves and axis.
Der vorliegende Beitrag versucht, einen Überblick über die aktuelle Zypern-Archäologie in Deutschland zu geben. Neben einer Zusammenfassung der Ausgrabungen und Feldforschungen, insbesondere seit dem 2. Weltkrieg, wird der Schwerpunkt auf den deutschen Sammlungen und Museen liegen, die zyprische Antiken beherbergen. Sie können an dieser Stelle nicht im Detail vorgestellt werden, doch soll durch die wichtigsten Eckdaten ihrer Entstehung und Zusammensetzung nicht nur ein Bild der Kypriaka in öffentlichem Besitz, sondern auch ein Baustein zu einer Geschichte des Sammelns zyprischer Altertümer in Deutschland geliefert werden. Eine Zusammenstellung von zypern-relevanten Ausstellungen sowie von universitären Promotions- und Forschungsprojekten rundet die Übersicht ab.
Am 25. Februar 1789 teilt Friedrich Schiller seinem Freund Christian Gottfried Körner mit: „Das lyrische Fach, das Du mir anweisest, sehe ich eher für ein Exilium, als für eine eroberte Provinz an. Es ist das kleinlichste und auch das undankbarste unter allen. Zuweilen ein Gedicht lasse ich mir gefallen […].“ Diese unverblümte Selbstaussage lässt Schillers Verhältnis zur Lyrik in einem bedenklichen Licht erscheinen: Weder scheint er sich auf dem Feld der lyrischen Dichtung heimisch gefühlt zu haben, noch hat er offenbar die Ausarbeitung von Gedichten für sonderlich lohnenswert gehalten. Gemildert wird diese entschiedene Distanzierung einzig durch den Umstand, dass er zugesteht, wenigstens „zuweilen“ im lyrischen Fach tätig werden zu wollen. Ist daraus nun zu schließen, dass Schiller seine Gedichte nur als gelegentliche Nebenprodukte begriffen hat?
Bryophytes (mosses, liverworts and hornworts) are the most basal lineages among extant land plants. However, there is controversy regarding their monophyletic or paraphyletic origin. Moreover, the most basal group of bryophytes also remains questionable. Due to this it is a matter of debate which lineage of bryophytes acts as transition link between bryophytes and pteridophytes. In order to resolve these issues we conducted a bioinformatics analysis on available chloroplast genomes of 1 green alga, 5 bryophytes, 4 pteridophytes, and 6 gymnosperms. Our study is based on multiple genome analysis through whole proteome comparison. The higher number of orthologous identified in Anthoceros (a hornwort), in comparison to other bryophytes considered, against all pteridophytes strongly support hornworts as transition link between bryophytes and pteridophytes.
Wherever there is a person's right, there is a corresponding duty imposed upon that person to respect the rights of others. This co-existence of rights and duties may be explained better by the principle of reciprocity of rights and duties. Such is the basis of Land as a Human Right: A History of Land Law and Practice in Tanzania. The esteemed author documents Tanzanian land law along its line of historical development (pre- and post-independence) whereby the thorny issues about 'rights' and 'duties' of the landed, landless and the intermediaries are elucidated. This volume is not limited to events in Tanzania, but includes jurisprudence of land law of other countries in order to tap some interpretative devices of our own by way of analogies. Various case types- reported and unreported, local and foreign- provide a tangible content to what would otherwise be pure theory. He also makes references to local newspapers as a way of tapping the public responses about land-related matters. His survey of such cases in and outside Tanzania led automatically to judgments touching on women's right to matrimonial property and inheritance; individual and collective rights to land; and the right to land of the indigenous peoples. It is the author's view that land law has remained poorly documented in Tanzania. There is plenty of literature about Land Law, yet these sources are not easily available or even accessible to every interested person. Equally, some of the available literature is so old that it may not always depict land law and/or practice as we tend to understand it today. This volume is a comprehensive text on land law in which all the necessary land law principles are highlighted with great precision. Advocate Rwegasira does this with a human rights approach, believing that it is through this approach that a person's right to land, whether individual or collective, can best be explained, especially in this era when conflict over land is unabatedly becoming central in family, communal and societal relations. The language of human rights is for all of us to speak. It follows, therefore, that practitioners both of the bar and the bench will also find it useful for quick reference, much as will do policy makers, law reformers and the general public in and outside Tanzania.

'Failure is not in my vocabulary' says Libertina Inaaviposa Amathila - medical doctor, leading member of Namibia's liberation movement SWAPO, and Cabinet Minister for 20 years. Insightful, candid and amusing, this book traces Libertina Amathila's journey from a village in western Namibia travelling alone to Dar es Salaam, Tanzania in 1962; medical training in Poland, Sweden and London; and the health and education centres in Zambia and Angola that she helped develop and run for Namibians in exile; to a victorious return home in 1989; service in the Cabinet of independent Namibia; and a leading role in the World Health Organisation. Courageous, committed, cutting through difficulties that deterred others, Libertina Amathila has assisted and empowered Namibian communities, particularly women, in exile and at home. As Minister of Regional and Local Government and Housing, Minister of Health and Social Services, and Deputy Prime Minister, she focused on those in need, such as squatters, street children, and those affected by HIV/AIDS, and undertook immediate practical measures to improve their lives. Packing her tent and supplies, she drove to remote areas and camped out until houses and clinics were built for marginalized communities, assisting in the design and construction process herself. An indomitable spirit drives this remarkable woman. This is her story.

One of the greatest challenges faced by African women writers is finding the time and the space to write. In November 2011 the third FEMRITE African Women Writers Residency alleviated this challenge for 15 women writers from 11 different countries across the continent. The writers from Kenya, Uganda, Malawi, Zimbabwe, Namibia, Tunisia, Ghana, Nigeria, South Africa, Cameroon, and Botswana gathered in Kampala, Uganda for two weeks. This anthology is the product of the writing developed before and during the residency. The short stories included are told from different perspectives, with varied voices, some experienced, others less so, but all told with freshness and honesty.

This book thematically tackles issues that relate to the perpetual struggle between the forces of control and the forces of mental and intellectual liberation in Africa and Kenya in particular. The book addresses the colonial legacy of poverty creation, as well as the socio-political conditioning of Africans to dislike each other and to be irresponsible and disunited in the face of external threats. Poverty, hatred of other Africans, and excessive dependency on European powers can be traced to the policies adopted by colonial officials. Related to these issues, is post-colonial Kenya's attempts to addresses the political developments, the involvement of different types of media in those developments, Kenya's foreign policy, and the problem of political party transition. Ultimately, there are topical issues that continue to affect Kenya which include the question of coalition politics, the lessons of the 2002 elections, the media and corruption, parliament and foreign policy, and Africa's relations with the United States of America.
Cell phones and the Internet have been the recipients of in-depth research on their increased and rapid integration into everyday life and the innovative appropriations associated with them in many societies. The cell phone has attracted particular attention in its perceived abilities to both enhance and destruct social relationships. Our increased access to social media and to the cell phone has taken social networking to an unprecedented level. These communication technologies are revered by many as great, all-purpose, all-positive communication devices in spite of their flaws. They are overwhelmingly bestowed with agency and superiority. Too often, they are idolized with little regard to how they affect and are affected by their users on a personal level. The mutual shaping between technology and society is not adequately acknowledged. Technologies, in spite of the seemingly endless possibilities offered by their many functions, can quite literally be sterile and useless objects outside of conscious and tangible human effort. Cell phones and the Internet, though undoubtedly capable of providing myriad beneficial opportunities for their users, need at long last to be put in their place. This book is a contribution in that regard. Kindled by her own intimate history with her cell phone and a growing curiosity about ICTs in general, this book is a culmination of Crystal Powell's thoughts, reactions to and interpretations of some of the literature on these technologies. The book draws on and critically reviews contributions by some leading authors on the social shaping of ICTs and social media to offer a more nuanced and complex understanding of technology in relation to those who use and are used by it.

This book makes an important contribution to existing knowledge on the processes of reading and comprehension by identifying the various approaches and corresponding theories. The book is organized in various chapters that cumulatively lead to our entry into the three key areas. Chapter One provides important background to reading as a skill, explaining the hidden dynamics that avoid the process and outcome of reading. Chapter Two deals with comprehension and vocabulary, both very important aspects of the reading process, while Chapter Three focuses on the relationship between reading, remembering and perception. Chapters four and five deal with various ways of assessing comprehension and the role of the reader respectively.
The study offers research into the efficacy of HIV and AIDS communication strategies for adolescents, especially with regards to selected secondary schools in Kenya. The study is a useful point of reference to both Kenyan researchers into HIV and AIDS as well as international scholars exploring Africanist perspectives of the socio-cultural dimensions of the pandemic.
In der vorliegenden prospektiven Studie in der neurochirurgischen Klinik an der Johann Wolfgang Goethe-Universität in Frankfurt am Main wurden Oktober 2001 bis August 2006 dreiunddreißig Patienten mit intraduralen Raumforderungen der zervikalen und thorakalen Wirbelsäule unter Betrachtung eines möglichen prädiktiven Wertes der Serumkonzentration des Gliaproteins S100B und des intraoperativen Monitorings untersucht.
Bei jedem Patienten erfolgte das intraoperative Monitoring mittels MEPs und SEPs, außerdem wurde prä- und mehrfach postoperativ die S100B-Serumkonzentration bestimmt. Anschließend wurden die Patienten mindestens 1 Jahr nach dem Eingriff in Bezug auf die noch bestehenden körperlichen Folgen sowie auch die Einschränkungen im alltäglichen Leben und im psychischen Wohlbefinden untersucht.
Der Zusammenhang zwischen dem neurologischen Langzeitoutcome, den Teilergebnissen im intraoperativen Monitoring und im postoperativen S100B-Verlauf zeigte sich deutlich und war statistisch signifikant. Die gemeinsame Betrachtung der Serumkonzentration des Gliaproteins und des IOM unterstrich diese Ergebnisse nochmals, die Spezifität für ein gutes Outcome bei unkompliziertem Monitoring und ausbleibenden Anstieg des S100B lag zum Beispiel bei 100%.
Auch in der Befragung zum alltäglichen Leben anhand des SF-36 zeigte sich ein klarer Zusammenhang zwischen dem klinischen Outcome und den subjektiv aus Patientensicht geschilderten Problemen in der Bewältigung des Alltags. Interessanterweise korrelierte aber die psychische Summenskala des Scores nicht mit dem körperlichen Befund. Die Gruppe des schlechten Outcomes zeigte ein besseres psychisches Wohlbefinden als die Gruppe des guten Outcomes, die aber mit der Normstichprobe vergleichbare Ergebnisse erzielte.
We have analysed the microseismic activity within the Rwenzori Mountains area in the western branch of the East African Rift. Seismogram recordings from a temporary array of up to 27 stations reveal approximately 800 events per month with local magnitudes ranging from –0.5 to 5.1. The earthquake distribution is highly heterogeneous. The majority of located events lie within faults zones to the East and West of the Rwenzoris with the highest seismic activity observed in the northeastern area, where the mountains are in contact with the rift shoulders. The hypocentral depth distribution exhibits a pronounced peak of seismic energy release at 15 km depth. The maximum extent of seismicity ranges from 20 to 32 km and correlates well with Moho depths that were derived from teleseismic receiver functions. We observe two general features: (i) beneath the rift shoulders seismicity extends from the surface down to ca. 30 km depth; (ii) beneath the rift valley seismicity is confined to depths greater than 10 km. From the observations there is no indication for a crustal root beneath the Rwenzori Mountains. The magnitude frequency distribution reveals a b-value of 1.1, which is consistent with the hypothesis that part of the seismicity is caused by magmatic processes within the crust. Fault plane solutions of 304 events were derived from P-polarities and SV/P amplitude ratios. More than 70 % of the source mechanisms exhibit pure or predominantly normal faulting. T-axis trends are highly uniform and oriented WNW-ESE, which is perpendicular to the rift axis and in good agreement with kinematic rift models. At the northernmost part of the region we observe a rotation of the T-axis trends to NEN-SWS, which may be indicative of a local perturbation of the regional stress field.

The majority of located events lie within faults zones to the east and west of the Rwenzoris with the highest seismic activity observed in the northeastern area, where the mountains are in contact with the rift shoulders. The hypocentral depth distribution exhibits a pronounced peak of seismic energy release at 15 km depth. The maximum extent of seismicity ranges from 20 to 32 km and correlates well with Moho depths that were derived from teleseismic receiver functions. We observe two general features: (i) beneath the rift shoulders, seismicity extends from the surface down to ca. 30 km depth; (ii) beneath the rift valley, seismicity is confined to depths greater than 10 km. From the observations there is no indication for a crustal root beneath the Rwenzori Mountains. The magnitude frequency distribution reveals a b-value of 1.1, which is consistent with the hypothesis that part of the seismicity is caused by magmatic processes within the crust. Fault plane solutions of 304 events were derived from P-polarities and SV/P amplitude ratios. More than 70 % of the source mechanisms exhibit pure or predominantly normal faulting. T-axis trends are highly uniform and oriented WNW–ESE, which is perpendicular to the rift axis and in good agreement with kinematic rift models. At the northernmost part of the region we observe a rotation of the T-axis trends to NEN–SWS, which may be indicative of a local perturbation of the regional stress field.

A new species of Culcua Walker (Diptera: Stratiomyidae), C. lingafelteri Woodley, new species, is described from northern Vietnam. It is diagnosed relative to other species using the recent revision of the genus by Rozkošný and Kozánek (2007). This is the first species of Culcua reported from Vietnam.
Since the mid-1980s, there has been much federalism talk in Cameroon where federation (said to have been created in Foumban in 1961) had supposedly been 'overwhelmingly' rejected in 1972 by Cameroonians. 'Confusioncracy' is the one good term that could conveniently explain it. Written with the trilogy of criticism, provocation, and construction in mind, this book aims at reconstructing a new and vigorous society in Cameroon that ensures respect for fundamental human rights and certain basic shared values. Much as the book centres on the Anglophone Problem; it is principally about human rights and their excessive violations - the direct result of the absence of separation of powers and constitutionalism. It largely condemns Cameroon's government for incessantly singing democracy and rule of law at the same time as it is massively torturing and wantonly killing citizens that dare to question the confusion. While sharing the position that a state like Cameroon must be seen to ensure that its laws and other practices accord with its international commitments, the book nonetheless strives to apportion the blame for Cameroon's human rights catastrophe accordingly; showing how the English-speaking minority itself, generally speaking, contributes to a large extent in propping up the dictatorship that is oppressing not only that minority but Cameroonians at large. The book challenges Cameroon to assume a leadership role in uniting Africans through meaningful federalization rather than further splitting them into incapable mini-states on the challenging world stage.
This is a complex volume that combines a good deal of survey data on Bakassi and its populations with more ethnographically based insights into the conditions of the Bakassi communities. The book is the outcome of research carried out by Fongot Kini between 2004 and 2009. The work is intended to serve as first hand exhaustive information on the live situation in the contested Bakassi Cameroon-Nigeria border region. The term Bakassi engenders multiple meanings loaded with many conflicting emotional, spiritual and material interests. Native inhabitants are systematically disinherited of their ancestral cultural heritage and socio-economic resources. They are bastardised, humiliated and scammed by unscrupulous opportunists who deliberately misidentify them with intentions of dispossessing them of their ancestral lands and natural resources. Overall the author is in sympathy with the Bakassi who he argues have been marginalised and neglected by the Cameroon state. In particular, the value of the indigenous communities in terms of local economies as well as securing this vital border area has not been recognised and various external groups have been either allowed or encouraged to settle there to both the detriment of local populations and to the security of the region.
The late Julius Kambarage Nyerere was nicknamed 'Musa' (Moses) during the later, post-independence years for leading his people from slavery and guiding them toward a free land of prosperity - the Promised Land. The Tanzanian odyssey chronicled in this book, which first appeared ten years ago as Tanzanians to the Promised Land, has been updated with new research. The author- also an engineer and a journalist- offers an enlightened and unbiased discussion of the journey and both sides of the contributions - successes and failures - made by former presidents and their systems of administration: the late Mwalimu Julius K. Nyerere, Alhajj Ali H. Mwinyi, and Mr. Benjamin W. Mkapa. Tanzanians' hopes and expectations of the incumbent president, H.E. Mr. Jakaya Mrisho Kikwete, are also discussed. It is not intended as a political campaign of any kind, for any party or any individual. As a brief, yet comprehensive guide to the understanding of our nation's political and economic history, it puts forward suggestions concerning important areas of the country's economic development. Nyerere unfortunately didn't live to see his people arrive at the hoped-for destination, and I. J. Werrema's original inspiration to write, at forty years of independence, is sustained because after fifty years The Promised Land is Still Too Far.

Nancy slaps palms with her friends and laughs a lot. She wears bell-bottom pants which swing when she walks through Uhuru Gardens. Nancy will finish secondary school this year, but she doesn't really know what will happen to her after that. Deo reads seriously, but he also spends many evenings in bars. He works in a factory laboratory, where his Form VI education elevates him above the other workers. He knows that there are some 'big men' who live off the sweat of the others at the factory; it isn't right, but what does a lone youth do about it? Deo also wants to marry Nancy. Magege, the manager of 'Mountain Goat Rubber Factory', has the means to fulfill all his personal wants-including his taste for young girls. Nancy's mother, Maria, has no private means except selling her own body and her dream of a better life for her daughter. The Wicked Walk swirls around the lives of these four, set on a backdrop of workers' struggles and the rhythm of Dar es Salaam as city dwellers, and especially youths, know it. In this searingly honest, and at times poignant, novel the author raises important questions about the position of women in society, the causes of prostitution, corrupt and inefficient managers, and the groupings of youth who struggle towards ideological clarity as they attempt to understand their society.
Aspects of Colonial Tanzanian History is a collection of essays that examines the lives and experiences of both colonizers and the colonized during colonial rule in what is today known as Tanzania. Dr. Mbogoni examines a range of topics hitherto unexplored by scholars of Tanzania history, namely: excessive alcohol consumption (the sundowners); adultery and violence among the colonial officials; attitudes to inter-racial sexual liaisons especially between Europeans and Africans; game-poaching; European settler vigilantism; radio broadcasting; film production and the nature of Arab slavery in Zanzibar. A particularly noteworthy case related to European vigilantism is examined: the trial of Oldus Elishira, a Maasai, for the murder of a European settler farmer in 1955. The victim, Harold M. Stuchbery, was speared to death when he attempted to 'arrest' a group of Maasai young men who were passing through his farm. The event highlighted the differences in the concepts of justice held by Maasai and the imported justice systems from the colonizers. It also raised vexing questions about the colonial judge's acquittal of Oldus Elishira, while the Maasai who should have been satisfied with that decision decided to take it upon themselves to mete out an appropriate punishment to Elshira instead of total acquittal, and to compensate Mrs. Stuchbery for the death of her husband by giving her a number of heads of cattle.

Anthony Mugo's Never Say Never is a compelling story of a teenager's quest for education under the most difficult conditions. Daniel Muthini Njoki, the son of a poor, single mother, is arrested and taken to a remand home in Murang'a, then to Getathuru Reception Centre. He is subsequently transferred to other approved schools: Kericho, Othaya, and finally Kabete, where he sits and passes the Kenya Certificate of Secondary Education. The doors to a university are now open. Although he is an innocent inmate, and although textual evidence points in the direction of the mother, the question of who engineered his arrest is part of what makes this work so unputdownable. The sum total is a superlatively well-written novel about the difficulties, the challenges, and the hopes of getting an education in Kenya.

This book explores the customary, social, economic political and rights issues surrounding access, ownership and control over land from a gender perspective. It combines theory and practice from researchers, lawyers and judges, each with track records of working on women and rights concerns. The nexus between the reluctance to recognize and materialize women?s right to land, and the increasing feminization of poverty is undeniable. The problem assumes special acuity in an essentially agrarian context like Cameroon, where the problem is not so much the law as its manner of application. That this book delves into investigating the principal sources and reasons for this prevalent injustice is particularly welcome. As some of the analyses reveal, denying women their right to land acquisition or inheritance is sometimes contrary to established judicial precedents and even in total dissonance with the country?s constitution. Traditional and cultural shibboleths associated with land acquisition and ownership that tend to stymie women?s development and fulfilment, must be quickly shirked, for such retrograde excuses can no longer find comfort in the law, morality nor in ?modern? traditional thinking. The trend, albeit timid, of appointing women to Land Consultative Boards and even as traditional authorities, can only be salutary. These are some positive practical steps that can translate the notion of equal rights into ?equal power? over land for both sexes; otherwise ?equality? in this context will remain an unattractive slogan.
The subject of revolutionary overthrow of constitutional orders in Africa is at the intersection of three disciplines: jurisprudence and legal philosophy, constitutional law and power politics, and civil-military relations, that is, military security policy which is one aspect of national security policy. The subject is of interest in at least four ways. It problematizes the inescapable question of governance in the African continent. It challenges the democratization agenda in Africa - how does one democratize not only political governance but also the instruments of violence in the state? It also challenges African constitutional lawyers and policy makers to seek a constitutional model that addresses the enduring menace of the power of the gun in African affairs and the changing role of the military in African politics. Finally, it underscores concerns about sovereignty and national security. This book contributes to a fuller understanding of the coup syndrome in African. To this end, it vigorously interrogates the place of coups in the governance of Africa, and explores the relevance of Kelsen's theory of revolutionary legality in the context of coup d'états in Africa. The Cameroon Condition brings together three seminal essays by George Ngwane, one of the most renowned, committed and daring Anglophone Cameroon writers. 'The Mungo Bridge, ' is a stinging indictment of the tenuous relations between La Republique du Cameroun and the Southern Cameroons - a marriage gone sour right from the honeymoon. It raises hard questions on the failed union, and is uncompromisingly courageous in the solutions it proposes. This popular essay was first published at a time when it was risky to be open and critical, especially on what has come to be known as The Anglophone Problem. 'The Anglophone File' discusses the narrow and barren politics of belonging that have exacerbated divisions and controversies among Anglophone elites, turning them into political fodder for the Francophone dominated state. The essay suggests ways out of the divisions and intrigue that have kept Anglophones permanently at daggers drawn against each other, and facilitated their exploitation, humiliation and marginalization. The third essay, 'Fragments of Unity, ' concerns the South West Region, whose leaders Ngwane criticizes of political opportunism and of a chronic lack of vision and fortitude with regard to the socio-economic development of the region. It calls for a leadership free of the docility, mediocrity and praise-singerliness. These are powerful essays that have attracted praise and criticism alike. They are essays to leave few indifferent. Their continued relevance to current debates makes of them a most read.

From its very inception, detective fiction has enjoyed a great popularity among the young and the old, the learned and the not so learned. By some unfortunate stroke of irony, its respect has not kept pace with its enormous popularity. For over half a century now, it has remained the bane of creative writing. In strict intellectual circles, it is very rare to find people talk defensively and interestingly about the genre. Yet Asong has chosen to do just that. He has stoutly defended the weak by putting up a good case for its continued existence. He has also shown how irresistible key elements of the genre are to even the best respected novelists. Finally he has demonstrated for the first time, how the genre has been domesticated by African writers of very great repute such as Ngugi, Sembene and Lessing. That he has been able to prove that these writers have used techniques of detective fiction is a significant broadening of the horizons for appreciating creative writing in Africa.
