Molecular cause and functional impact of altered synaptic lipid signaling due to a prg‐1 gene SNP

Loss of plasticity-related gene 1 (PRG-1), which regulates synaptic phospholipid signaling, leads to hyperexcitability via increased glutamate release altering excitation/inhibition (E/I) balance in cortical networks. A recently reported SNP in prg-1 (R345T/mutPRG-1) affects ~5 million European and US citizens in a monoallelic variant. Our studies show that this mutation leads to a loss-of-PRG-1 function at the synapse due to its inability to control lysophosphatidic acid (LPA) levels via a cellular uptake mechanism which appears to depend on proper glycosylation altered by this SNP. PRG-1(+/-) mice, which are animal correlates of human PRG-1(+/mut) carriers, showed an altered cortical network function and stress-related behavioral changes indicating altered resilience against psychiatric disorders. These could be reversed by modulation of phospholipid signaling via pharmacological inhibition of the LPA-synthesizing molecule autotaxin. In line, EEG recordings in a human population-based cohort revealed an E/I balance shift in monoallelic mutPRG-1 carriers and an impaired sensory gating, which is regarded as an endophenotype of stress-related mental disorders. Intervention into bioactive lipid signaling is thus a promising strategy to interfere with glutamate-dependent symptoms in psychiatric diseases.
Left ventricular non-compaction cardiomyopathy and left ventricular assist device: a word of caution

BACKGROUND: In patients with left ventricular non-compaction (LVNC), implantation of a left ventricular assist device (LVAD) may be performed as a bridge to transplantation. In this respect, the particular characteristics of the left ventricular myocardium may represent a challenge.
CASE PRESENTATION: We report a patient with LVNC who required urgent heart transplantation for inflow cannula obstruction nine months after receiving a LVAD. LVAD parameters, echocardiography and examination of the explanted heart suggested changes of left ventricular configuration brought about by LVAD support as the most likely cause of inflow cannula obstruction.
CONCLUSIONS: We conclude that changes experienced by non-compacted myocardium during LVAD support may give rise to inflow cannula obstruction and flow reduction. Presence of LVNC mandates tight surveillance for changes in LV configuration and LVAD flow characteristics and may justify urgent transplantation listing status.
The aim of this clinical trial was to evaluate the impact of all-trans retinoic acid (ATRA) in combination with chemotherapy and to assess the NPM1 status as biomarker for ATRA therapy in younger adult patients (18-60 years) with acute myeloid leukemia (AML). Patients were randomized for intensive chemotherapy with or without open-label ATRA (45 mg/m2, days 6-8; 15 mg/m2, days 9-21). Two cycles of induction therapy were followed by risk-adapted consolidation with high-dose cytarabine or allogeneic hematopoietic cell transplantation. Due to the open label character of the study, analysis was performed on an intention-to-treat (ITT) and a per-protocol (PP) basis. One thousand one hundred patients were randomized (556, STANDARD; 544, ATRA) with 38 patients treated vice versa. Median follow-up for survival was 5.2 years. ITT analyses revealed no difference between ATRA and STANDARD for the total cohort and for the subset of NPM1-mutated AML with respect to event-free (EFS; p = 0.93, p = 0.17) and overall survival (OS; p = 0.24 and p = 0.32, respectively). Pre-specified PP analyses revealed better EFS in NPM1-mutated AML (p = 0.05) and better OS in the total cohort (p = 0.03). Explorative subgroup analyses on an ITT basis revealed better OS (p = 0.05) in ATRA for genetic low-risk patients according to ELN recommendations. The clinical trial is registered at clinicaltrialsregister.eu (EudraCT Number: 2004-004321-95).
Cl(-) plays a crucial role in neuronal function and synaptic inhibition. However, the impact of neuronal morphology on the diffusion and redistribution of intracellular Cl(-) is not well understood. The role of spines in Cl(-) diffusion along dendritic trees has not been addressed so far. Because measuring fast and spatially restricted Cl(-) changes within dendrites is not yet technically possible, we used computational approaches to predict the effects of spines on Cl(-) dynamics in morphologically complex dendrites. In all morphologies tested, including dendrites imaged by super-resolution STED microscopy in live brain tissue, spines slowed down longitudinal Cl(-) diffusion along dendrites. This effect was robust and could be observed in both deterministic as well as stochastic simulations. Cl(-) extrusion altered Cl(-) diffusion to a much lesser extent than the presence of spines. The spine-dependent slowing of Cl(-) diffusion affected the amount and spatial spread of changes in the GABA reversal potential thereby altering homosynaptic as well as heterosynaptic short-term ionic plasticity at GABAergic synapses in dendrites. Altogether, our results suggest a fundamental role of dendritic spines in shaping Cl(-) diffusion, which could be of relevance in the context of pathological conditions where spine densities and neural excitability are perturbed.
Der jeweilige Betrugsbegriff kristallisiert sich in der Interpretation des § 263 StGB. Obwohl die Vorschrift seit 1875 existiert, hat sich ihre Interpretation von Epoche zu Epoche und in einer Reihe von Aspekten geändert. Die sich so entwickelten Betrugsbegriffe sind nicht frei von Vagheit und Widersprüchen.
Mittels einer Analyse der Rechtsprechung des Reichsgerichts und des Bundesgerichtshofs zwischen 1879 und 1979 arbeite ich heraus, welche ökonomischen und gesellschaftlichen Entwicklungen und Tendenzen die sich verändernden Betrugsbegriffe bestimmen. Dabei werden nicht nur juristische Argumente analysiert, die Urteilsgründe, sondern auch die Sachverhaltsdarstellungen mit Blick auf die gesellschaftlichen und ökonomischen Positionen der Beteiligten und Betroffenen und ihrer Beziehungen. So können auch latente Aspekte der Begriffe aufgefunden und explizit gemacht werden. Es wird gezeigt, dass einige Formen von Täuschung und Manipulation, im wesentlichen durch Werbung, aus dem Betrugsbegriff ausgenommen werden ebenso wie einige Formen von Vermögensbeschädigung, während auf der anderen Seite Schäden einbezogen werden, die nicht wirklich Vermögensschäden sind
Es wird auch gezeigt, dass der Wandel des politischen Regimes die Rechtsprechung und ihren Betrugsbegriff beeinflusst.
Die Analyse wird mittels qualitativer und quantitativer Inhaltsanalyse vorgenommen. Außerdem entwickle ich einen Betrugsbegriff der mit einer konstitutionellen Demokratie eher vereinbar ist. Die Untersuchung wurde zwischen 1981 und 1983 vorgenommen, das Buch 1985 veröffentlicht, was hier unverändert herausgegeben wird.
Wie wenige literarische Werke stehen die Romane von Bret Easton Ellis im Zeichen von Serie und Wiederholung. Sie bersten vor Motiv- und Form-Wiederholungen, sind mit ihren intertextuellen Zitaten einerseits Fortsetzungen voneinander, andererseits "Wiederholungen" von Figuren aus den Texten anderer Autoren und Produzenten, sie sind besessen von der Serialität der elektronischen Medien, und - last but not least - sie warten mit einem Serienmörder (in 'American Psycho') und mit Anschlagsserien (in 'Glamorama') auf. Damit überschreiten sie die analytische Unterscheidung der Bereiche des Seriellen, die Christine Blättler in ihrem enzyklopädischen Aufsatz über die Serie aufgestellt hat: Produktion, Präsentation und Narration sind bei Ellis gleichzeitig von Wiederholungen und Serien gekennzeichnet.
We investigate the effect of overreaction in the fine art market. Using a unique sample of auction prices of modern prints, we define an overvalued (undervalued) print as a print that was bought for a price above (below) its high (low) auction pricing estimate. Based on the overreaction hypothesis, we predict that overvalued (undervalued) prints generate a negative (positive) excess return at a subsequent sale. Our empirical findings confirm our expectations. We report that prints that were bought for a price 10 percent above (below) its high (low) pricing estimate generate a positive (negative) excess return of 12 percent (17 percent) after controlling for the general price movement on the prints market. The price correction for overvalued (undervalued) prints is more pronounced during recessions (expansions).
Paul Kammerer, der 1919 sein Buch 'Das Gesetz der Serie' publizierte - das wohl ehrgeizigste und umfangreichste Buch dieser Art, das je veröffentlicht wurde - gehört in die Reihe der leidenschaftlichen Sammler von Zufällen. Und er bestätigt die schon bei Wilhelm von Scholz bemerkte Neigung, die Welt der Zufälle zur Konkurrentin der Normalität kausalen Denkens zu machen und aus dem scheinbar Zufälligen eine Welt zu konstruieren, die von der Normalerfahrung verdeckt ist - eine Welt der ungestörten Bedeutsamkeit. Die Zufälle, die der Biologe Kammerer viele Jahre hindurch in seine Merkbücher eintrug, werden schließlich durch seine Theorie verwandelt in Erscheinungen der Serialität, von Wiederholung und Periodizität, wodurch das Tor aufgestoßen werde zu einer neuen Gesetzlichkeit. Wo Kausalität war, sollte Serialität werden. Das scheint der Sinn des Buchtitels 'Das Gesetz der Serie' zu sein, das in gewisser Weise ein Gegenprogramm zu Freuds: "Wo Es war, soll Ich werden" formuliert. Denn es soll eben nicht der 'subjektive' Zufall sein, die in die kausalgesetzliche Naturordnung eingewobene Zufälligkeit. Daneben soll vielmehr eine zweite Ordnung aufscheinen, die nicht weniger regelmäßig ist. Man muss sich vor Augen halten, dass Paul Kammerer nicht eigentlich ein Buch über, sondern eines gegen den Zufall geschrieben hat, über den er nur mit Herablassung oder Verachtung spricht. Von Zufall dürfe, erklärt er, keinesfalls geredet werden: "Ganz abgesehen davon, dass die Erklärung einer Begebenheit durch 'Zufall' ein grober Missbrauch ist und in keiner wissenschaftlichen Begründung geduldet werden sollte." Der Zufall dient ihm nur dazu, das Tor zum Gebiet seiner Serialitätsforschung abzustecken, das er mit keinem geringerem wissenschaftlichen Ernst ins Auge fasst als Sigmund Freud die Träume und pathologischen Phänomene des Alltags.
In Menschenobhut können junge von Menschenhand aufgezogene Dachse sehr zahm werden. Daher kennt man eine Eigenheit der Dachsheit, auf die es mir hier ankommt: Man kann Dachse nur bedingt erziehen. Sie reagieren nämlich nicht auf negative Verstärker, wie man im Fachjargon all jene Bestrafungstechniken von Ausschimpfen, Futterentzug über Prügel bis zu Elektroschocks nennt, die immer noch die Grundlage fast jeder Dressur, sei es im Zirkus, im Pferdesport oder Hundespektakel bilden. Der Verhaltensforscher Irenäus Eibl-Eibesfeldt ist an dieser Eigenheit des Dachses im Falle seines von Hand aufgezogenen zahmen Dachses schier verzweifelt, bis er erkannte: Dass es Tiere gibt, die die Reaktion auf den negativen Verstärker nicht in ihrem Verhaltensprogramm haben und somit undressierbar sind. Eibl mochte seinen Dachs aber trotzdem, und nachdem er es aufgegeben hatte, dem Dachs das freie Geruchsmarkieren im Haus verbieten zu wollen, gestaltete sich das Zusammenleben auch wieder ohne Verzweiflung. Damit bin ich bei meinem Titel angekommen: Reden wie ein Hund - Pfeifen wie eine Maus - Lernen wie ein Dachs. Über abgebrochene Serien des Tier-Werdens in Kafkas Erzählungen. Das Vorhaben ist der Versuch, anhand dreier Erzählungen Kafkas - nämlich die 'Forschungen eines Hundes', 'Josefine, die Sängerin' und 'Der Bau' - eine Art Entwicklungslinie im Tier-Werden Kafkas aufzuzeigen, die mit dem unerziehbaren Dachs abbricht, aber damit nicht, wie man Kafka oft unterstellt hat, im unentschiedenen Nichts landet, sondern einen Ausweg zeigt, mit dem man leben kann. Das hoffe ich zum Ende deutlich machen zu können.
Dezember 1925, Staatsoper Unter den Linden Berlin: Zwei Tage vor Weihnachten besuchen Theodor W. Adorno und Walter Benjamin die erste auf die aufsehenerregende Uraufführung folgende Vorstellung von Alban Bergs Oper Wozzeck: Es dirigiert Erich Kleiber, die Hauptpartien singen Leo Schützendorf, Fritz Soot und Sigrid Johanson. Nicht nur auf den 'Expertenhörer' Adorno - der in einem an seinen Kompositionslehrer übermittelten Bericht sowohl Benjamins Bestimmung des "Ausdruckslosen" aufgreift, als auch dessen physische Präsenz erwähnt - hinterlässt die Musik einen bleibenden Eindruck. Benjamin erinnert sich dieser Aufführung noch zehn Jahre später in einem seiner Briefe an Adorno und bekennt nach der Lektüre von dessen Berg-Analysen, "dass der überwältigende Eindruck, kraft dessen mich der Wozzeck an jenem Abend gefangen nahm, das Signal eines mir unbewussten aber bis ins einzelne nennbaren Betroffenseins gewesen ist". Von der geschichtsphilosophischen Einordnung der Berg'schen Musik, welche "wie die gleichsam namenlose technische Arbeit des Schülers Schönbergs die Tradition des 19ten Jahrhunderts im Namen des Musikers zur Ruhe geleitet und ihren eigenen Klagegesang anstimmen lässt", führt Benjamin sich dann sogar versucht, den "Begriff des 'kleinen Übergangs' [...] der Kompositionslehre abzuborgen (GB V, 565). Im Gegensatz zu Adorno wird Walter Benjamins Theorie jedoch kaum mit Musik in Verbindung gebracht, sondern vor allem für ihr Bilddenken geschätzt. Auch wenn im 'Ursprung des Deutschen Trauerspiels', in den Erinnerungsszenen der 'Berliner Kindheit', den Häuser- und Gedankenschluchten der 'Passagenarbeit' und nicht zuletzt in seinem Briefwechsel durchaus von Musik die Rede ist, rücken Klänge seltener in den Mittelpunkt seiner Schriften und sind in der Rezeption bisher auch weniger beachtet worden. Ohnedies wird derjenige enttäuscht, der gerade von Benjamin - so reizvoll es sein mag, sich solchen Gegenständen widmende Essays zu imaginieren - die Exegese des gängigen musikalischen Kanons erwartet: Keine Abhandlung zu Schuberts Streichquintett schließt an die Interpretation der 'Wahlverwandschaften' an und ebenso vergeblich sucht man nach die Erwägungen zum Trauerspiel auf die Affektkontrolle der 'opera seria' oder die des Kraus-Essays auf die Musik des 'Fackel'-Habitués Arnold Schönberg übertragenden Arbeiten. Dennoch kommt Musik, Klage und Tönen in Benjamins Reflexionen eine nicht unerhebliche Bedeutung zu. Selten drängen sie sich lautstark in den Vordergrund, erlauben aber - zumal in einer Zeit, in der auch die professionelle Auseinandersetzung mit Musik die ideale Beständigkeit musikalischer Werke und ihrer Texte zunehmend in Zweifel zieht - theoretische Einsichten, über die das auf andere Dinge gerichtete Ohr des Experten achtlos hinweggeht. In Ergänzung zur wohlbekannten epistemischen und poetischen Rolle seiner Sprach- und Denkbilder unternimmt dieser Band den ersten systematisch angelegten Versuch, sowohl den kultur- und medienhistorischen Zusammenhängen von Benjamins akustischen Motiven nachzugehen als auch ihre ästhetische Relevanz für die gegenwärtige Produktion und Reproduktion von Klängen zu reflektieren.
'The Making of Americans' wurde 1903 begonnen, 1905/6 zugunsten des Erzählbands 'Three Lives' unterbrochen, 1906 und 1908 komplett umgeschrieben und 1911 schließlich abgeschlossen (also viel früher als 'Ulysses' und 'À la recherche du temps perdu'). Zur Veröffentlichung des Romans kam es allerdings erst 1925, was die öffentliche Wahrnehmung des Textes als einer Urszene der Moderne stark beeinträchtigt hat. Aber es gibt auch noch andere Gründe, weshalb 'The Making of Americans' rasch in Vergessenheit geriet: der riesige Umfang des Romans (fast 1000 Seiten) und seine serielle Wiederholungsstruktur sorgten dafür, dass er einer größeren Leseöffentlichkeit bis heute weitgehend unbekannt blieb. Steins Aufkündigung der Erzähltradition des 19. Jahrhunderts fiel noch radikaler aus als Prousts und Joyces Romanexperimente. Es gibt aber durchaus Gemeinsamkeiten: alle drei Autoren machen die Prozesse des Erinnerns, Denkens und Fühlens zur Grundlage der Darstellung im Roman und verzichten auf die teleologische Konsequenz kausaler Verknüpfungen zugunsten des verdichteten Augenblicks. Schon im ursprünglichen Entwurf von 'The Making of Americans', der auf den ersten Seiten des Romans noch erkennbar ist, sind erste Anzeichen für den Bruch mit der Erzählform erkennbar. Stein wollte Amerika als neue Nation schildern, "whose tradition it had taken scarcely sixty years to create". Im Mittelpunkt sollten drei Generationen zweier Immigrantenfamilien stehen, die Dehnings und die Herslands. "The old people in a new world, the new people made out of old, that is the story that I mean to tell, for that is what really is and what I really know." Obwohl sie den Roman als Gesellschaftspanorama anlegte, konzentrierte sich Stein von Anfang an auf das Privatleben ihrer Protagonisten. Im Gegensatz zu den Realisten des 19. Jahrhunderts, die den Konflikt von Neigung und Pflicht, von sozialen Normen und subjektiver Moral in den Mittelpunkt der Handlung stellten, folgte Stein dem Programm des Naturalismus und richtete ihr Interesse nicht auf die Handlungen und Handlungsspielräume der Subjekte, sondern auf die gesellschaftlichen Kräfte, die Subjektivität formen und prägen. Der Titel 'The Making of Americans' fasst die Ausbildung amerikanischer Identität als einen beinah industriellen Herstellungsprozess sozialer Identität und deutet darin ein erstes Motiv für eine serielle Schreibweise an.
Desert dust is one of the most abundant ice nucleating particle types in the atmosphere. Traditionally, clay minerals were assumed to determine the ice nucleation ability of desert dust and constituted the focus of ice nucleation studies over several decades. Recently some feldspar species were identified to be ice active at much higher temperatures than clay minerals, redirecting studies to investigate the contribution of feldspar to ice nucleation on desert dust. However, so far no study has shown the atmospheric relevance of this mineral phase.
For this study four dust samples were collected after airborne transport in the troposphere from the Sahara to different locations (Crete, the Peloponnese, Canary Islands, and the Sinai Peninsula). Additionally, 11 dust samples were collected from the surface from nine of the biggest deserts worldwide. The samples were used to study the ice nucleation behavior specific to different desert dusts. Furthermore, we investigated how representative surface-collected dust is for the atmosphere by comparing to the ice nucleation activity of the airborne samples. We used the IMCA-ZINC setup to form droplets on single aerosol particles which were subsequently exposed to temperatures between 233 and 250 K. Dust particles were collected in parallel on filters for offline cold-stage ice nucleation experiments at 253–263 K. To help the interpretation of the ice nucleation experiments the mineralogical composition of the dusts was investigated. We find that a higher ice nucleation activity in a given sample at 253 K can be attributed to the K-feldspar content present in this sample, whereas at temperatures between 238 and 245 K it is attributed to the sum of feldspar and quartz content present. A high clay content, in contrast, is associated with lower ice nucleation activity. This confirms the importance of feldspar above 250 K and the role of quartz and feldspars determining the ice nucleation activities at lower temperatures as found by earlier studies for monomineral dusts. The airborne samples show on average a lower ice nucleation activity than the surface-collected ones. Furthermore, we find that under certain conditions milling can lead to a decrease in the ice nucleation ability of polymineral samples due to the different hardness and cleavage of individual mineral phases causing an increase of minerals with low ice nucleation ability in the atmospherically relevant size fraction. Comparison of our data set to an existing desert dust parameterization confirms its applicability for climate models. Our results suggest that for an improved prediction of the ice nucleation ability of desert dust in the atmosphere, the modeling of emission and atmospheric transport of the feldspar and quartz mineral phases would be key, while other minerals are only of minor importance.

In situ single particle analysis of ice particle residuals (IPRs) and out-of-cloud aerosol particles was conducted by means of laser ablation mass spectrometry during the intensive INUIT-JFJ/CLACE campaign at the high alpine research station Jungfraujoch (3580 m a.s.l.) in January–February 2013. During the 4-week campaign more than 70 000 out-of-cloud aerosol particles and 595 IPRs were analyzed covering a particle size diameter range from 100 nm to 3 µm. The IPRs were sampled during 273 h while the station was covered by mixed-phase clouds at ambient temperatures between −27 and −6 °C. The identification of particle types is based on laboratory studies of different types of biological, mineral and anthropogenic aerosol particles. The outcome of these laboratory studies was characteristic marker peaks for each investigated particle type. These marker peaks were applied to the field data. In the sampled IPRs we identified a larger number fraction of primary aerosol particles, like soil dust (13 ± 5 %) and minerals (11 ± 5 %), in comparison to out-of-cloud aerosol particles (2.4 ± 0.4 and 0.4 ± 0.1 %, respectively). Additionally, anthropogenic aerosol particles, such as particles from industrial emissions and lead-containing particles, were found to be more abundant in the IPRs than in the out-of-cloud aerosol. In the out-of-cloud aerosol we identified a large fraction of aged particles (31 ± 5 %), including organic material and secondary inorganics, whereas this particle type was much less abundant (2.7 ± 1.3 %) in the IPRs. In a selected subset of the data where a direct comparison between out-of-cloud aerosol particles and IPRs in air masses with similar origin was possible, a pronounced enhancement of biological particles was found in the IPRs.
In-situ single particle analysis of ice particle residuals (IPR) and out-of-cloud aerosol particles was conducted by means of laser ablation mass spectrometry during the intensive INUIT-JFJ/CLACE campaign at the high alpine research station Jungfraujoch (3580 m a.s.l.) in January/February 2013. During the four week campaign more than 70000 out-of-cloud aerosol particles and 595 IPR were analyzed covering a particle size diameter range from 100 nm to 3 μm. The IPR were sampled during 273 hours while the station was covered by mixed-phase clouds at ambient temperatures between -27 °C and -6 °C. The identification of particle types is based on laboratory studies of different types of biological, mineral and anthropogenic aerosol particles. As outcome instrument specific marker peaks for the different investigated particle types were obtained and applied to the field data. The results show that the sampled IPR contain a larger relative amount of natural, primary aerosol, like soil dust (13 %) and minerals (11 %), in comparison to out-of-cloud aerosol particles (2 % and <1 %, respectively). Additionally, anthropogenic aerosol particles, like particles from industrial emissions and lead-containing particles, were found to be more abundant in the IPR than in the out-of-cloud aerosol. Size-resolved long-term measurements of atmospheric aerosol and cloud condensation nuclei (CCN) concentrations and hygroscopicity were conducted at the remote Amazon Tall Tower Observatory (ATTO) in the central Amazon Basin over a 1-year period and full seasonal cycle (March 2014–February 2015). The measurements provide a climatology of CCN properties characteristic of a remote central Amazonian rain forest site.
The CCN measurements were continuously cycled through 10 levels of supersaturation (S = 0.11 to 1.10 %) and span the aerosol particle size range from 20 to 245 nm. The mean critical diameters of CCN activation range from 43 nm at S = 1.10 % to 172 nm at S = 0.11 %. The particle hygroscopicity exhibits a pronounced size dependence with lower values for the Aitken mode (κAit = 0.14 ± 0.03), higher values for the accumulation mode (κAcc = 0.22 ± 0.05), and an overall mean value of κmean = 0.17 ± 0.06, consistent with high fractions of organic aerosol.
The hygroscopicity parameter, κ, exhibits remarkably little temporal variability: no pronounced diurnal cycles, only weak seasonal trends, and few short-term variations during long-range transport events. In contrast, the CCN number concentrations exhibit a pronounced seasonal cycle, tracking the pollution-related seasonality in total aerosol concentration. We find that the variability in the CCN concentrations in the central Amazon is mostly driven by aerosol particle number concentration and size distribution, while variations in aerosol hygroscopicity and chemical composition matter only during a few episodes.
For modeling purposes, we compare different approaches of predicting CCN number concentration and present a novel parametrization, which allows accurate CCN predictions based on a small set of input data.
