Orts- und zeitaufgelöste Elektronendichte eines gepulsten induktiv gekoppelten Entladungsplasmas

In der vorliegenden Bachelorarbeit wurde ein Modell für die räumlich und zeitlich aufgelöste Elektronendichteverteilung in einem gepulsten induktiv gekoppelten Plasma erstellt. Experimentell war es, bedingt durch den gepulsten Betrieb und die Wahl der Diagnostikmethode im Experiment „Prometheus“, nur möglich über die Zeit und den Ort gemittelte Elektronendichten zu messen.
Um nun den räumlichen Verlauf der Elektronendichte zu bestimmen, wurde die räumliche Elektronendichteverteilung durch eine ambipolare homogene Diffusion beschrieben. Die daraus resultierende Differentialgleichung wurde mithilfe von sphärischen Koordinaten unter Annahme von Azimutal- und Polarwinkelsymmetrie gelöst.
Der zeitliche Elektronendichteverlauf wurde durch die, für diesen Elektronendichtebereich gültige, Proportionalität zwischen elektrischer Leistung im Plasma und Elektronendichte berechnet. Die elektrische Leistung und deren zeitlicher Verlauf im Plasma ließ sich über ein Photodiodensignal im experimentellen Aufbau ermitteln.
Das so ermittelte Modell wurde auf die gemessenen integrierten Elektronendichten des Experiments „Prometheus“ angewendet. Durch das Modell ließ sich eine Aussage über die tatsächliche maximale Elektronendichte innerhalb des Entladungspulses treffen.
Protein turnover and quality control by the proteasome is of paramount importance for cell homeostasis. Dysfunction of the proteasome is associated with aging processes and human diseases such as neurodegeneration, cardiomyopathy, and cancer. The regulation, i.e. activation and inhibition of this fundamentally important protein degradation system, is still widely unexplored. We demonstrate here that the evolutionarily highly conserved type II triple-A ATPase VCP and the proteasome inhibitor PSMF1/PI31 interact directly, and antagonistically regulate proteasomal activity. Our data provide novel insights into the regulation of proteasomal activity.
In der vorliegenden Arbeit wurden Messungen zur Plasmadynamik eines Lorentz-Drift- Beschleunigers (LDB) durchgeführt. Dieser basiert auf einer koaxialen Elektrodengeometrie. Bei einem Überschlag führt der entstehende Stromfluss zu einemMagnetfeld, sodass die gebildeten Ladungsträger durch die resultierende Lorentzkraft beschleunigt werden. Es hat sich gezeigt, dass die Abhängigkeit von Durchbruchspannung und Druck dem charakteristischen Verlauf einer Paschenkurve folgt.
Die Strom-Spannungs-Charakteristik des Versuchsaufbaus wurde in Konfigurationen mit und ohne Funkenstrecke untersucht. Mit Hilfe von diesem als Schalter fungierenden Spark-Gaps konnte bei Durchbruchspannungen gemessen werden, die oberhalb des Selbstdurchbruchs liegen.
Es zeigte sich, dass die im Versuchaufbau verwendete Funkenstrecke keinen wesentlichen Einfluss auf die Entladung hat. Es kommt an der Funkenstrecke lediglich zu einem Spannungsabfall im Bereich einiger hundert Volt, der den Verlauf derEntladung im LDB allerdings nicht beeinflusst.
Der Lorentz-Drift-Beschleuniger könnte in Zukunft zur Erzeugung eines Druckgradienten verwendet werden, indem Teilchen von einem Rezipienten in einen Zweiten beschleunigt werden. Als Voruntersuchung zur Eingnung dieses als Lorentz-Drift-Ventil bezeichneten Konzeptes wurden Messungen durchgeführt, die den Einfluss der Durchbruchspannung auf die Teilchenbeschleunigung mit Hilfe eines piezokeramischen Elementes untersuchen. So wurde der magnetische Druck bzw. die entsprechende Kraft einer Entladungswolke in Abhängigkeit von Durchbruchspannungen bis etwa 9,5 kV untersucht. Es hat sich gezeigt, dass der Einsatz von hohen Spannungen sinnvoll ist, da sich die auf das Piezoelement einwirkende Kraft quadratisch zur Durchbruchspannung verhält. So wurde die maximale Kraft von 0,44N bei einer Zündspannung von 9,52 kV gemessen.
Zudem wurde untersucht, in welchem Druckbereich der Einfluss der Druckwelle zu messen und wie sich die Geschwindigkeit der Ausbreitung der Druckwelle bei verschiedenen Durchbruchspannungen verhält. Bei einer Entfernung von 231mm zwischen Elektrodengeometrie und Piezoelement hat sich gezeigt, dass im Druckbereich unterhalb von etwa 0,2mbar kein wesentlicher Einfluss des Gasdruckes auf die Piezospannung erkennbar ist. Dies lässt sich durch die geringe Teilchenanzahl im Arbeitsgas begründen, sodass Teilchenstöße vernachlässigt werden können. Die maximale gemessene Geschwindigkeit der durch die Entladung verursachten Druckwelle liegt bei 55 km s ± 10%.
Die gemessene Plasmadynamik lässt darauf schließen, dass das Konzept eines gepulsten Lorentz-Drift-Ventils insbesondere mit hohen Durchbruchspannungen realisierbar ist. Zur Erzeugung eines dauerhaften Druckgradienten müsste die Repetitionsrate allerdings ausreichend hoch sein, sodass der rückfließende Gasdurchsatz geringer ist als die durch den LDB erzeugte Drift. Geht man von der Schallgeschwindigkeit als Rückflussgeschwindigkeit der Teilchen aus, so sind mindestens Repetitionszeiten im Bereich einer Millisekunde erforderlich.
Ergänzend zu den durchgeführten Untersuchungen ist es sinnvoll, die bisherigen Messungen durch Einbau eines Triggers zu verifizieren. Ein Trigger erzeugt eine Vorentladung mit deren Hilfe die eigentliche Entladung auch im Bereich unterhalb des Selbstdurchbruchs gezündet werden kann.
The Fisher information constitutes a natural measure for the sensitivity of a probability distribution with respect to a set of parameters. An implementation of the stationarity principle for synaptic learning in terms of the Fisher information results in a Hebbian self-limiting learning rule for synaptic plasticity. In the present work, we study the dependence of the solutions to this rule in terms of the moments of the input probability distribution and find a preference for non-Gaussian directions, making it a suitable candidate for independent component analysis (ICA). We confirm in a numerical experiment that a neuron trained under these rules is able to find the independent components in the non-linear bars problem. The specific form of the plasticity rule depends on the transfer function used, becoming a simple cubic polynomial of the membrane potential for the case of the rescaled error function. The cubic learning rule is also an excellent approximation for other transfer functions, as the standard sigmoidal, and can be used to show analytically that the proposed plasticity rules are selective for directions in the space of presynaptic neural activities characterized by a negative excess kurtosis.
We present an effective model for timing-dependent synaptic plasticity (STDP) in terms of two interacting traces, corresponding to the fraction of activated NMDA receptors and the concentration in the dendritic spine of the postsynaptic neuron. This model intends to bridge the worlds of existing simplistic phenomenological rules and highly detailed models, thus constituting a practical tool for the study of the interplay of neural activity and synaptic plasticity in extended spiking neural networks. For isolated pairs of pre- and postsynaptic spikes, the standard pairwise STDP rule is reproduced, with appropriate parameters determining the respective weights and timescales for the causal and the anticausal contributions. The model contains otherwise only three free parameters, which can be adjusted to reproduce triplet nonlinearities in hippocampal culture and cortical slices. We also investigate the transition from time-dependent to rate-dependent plasticity occurring for both correlated and uncorrelated spike patterns.
Generating functionals may guide the evolution of a dynamical system and constitute a possible route for handling the complexity of neural networks as relevant for computational intelligence.We propose and explore a new objective function, which allows to obtain plasticity rules for the afferent synaptic weights. The adaption rules are Hebbian, self-limiting, and result from the minimization of the Fisher information with respect to the synaptic flux. We perform a series of simulations examining the behavior of the new learning rules in various circumstances.The vector of synaptic weights aligns with the principal direction of input activities, whenever one is present. A linear discrimination is performed when there are two or more principal directions; directions having bimodal firing-rate distributions, being characterized by a negative excess kurtosis, are preferred. We find robust performance and full homeostatic adaption of the synaptic weights results as a by-product of the synaptic flux minimization. This self-limiting behavior allows for stable online learning for arbitrary durations.The neuron acquires new information when the statistics of input activities is changed at a certain point of the simulation, showing however, a distinct resilience to unlearn previously acquired knowledge. Learning is fast when starting with randomly drawn synaptic weights and substantially slower when the synaptic weights are already fully adapted.
Polo-like kinase 1 regulates the stability of the mitotic centromere-associated kinesin in mitosis

Proper bi-orientation of chromosomes is critical for the accurate segregation of chromosomes in mitosis. A key regulator of this process is MCAK, the mitotic centromere-associated kinesin. During mitosis the activity and localization of MCAK are regulated by mitotic key kinases including Plk1 and Aurora B. We show here that S621 in the MCAK’s C-terminal domain is the major phosphorylation site for Plk1. This phosphorylation regulates MCAK’s stability and facilitates its recognition by the ubiquitin/proteasome dependent APC/CCdc20 pathway leading to its D-box dependent degradation in mitosis. While phosphorylation of S621 does not directly affect its microtubule depolymerising activity, loss of Plk1 phosphorylation on S621 indirectly enhances its depolymerization activity in vivo by stabilizing MCAK, leading to an increased level of protein. Interfering with phosphorylation at S621 causes spindle formation defects and chromosome misalignments. Therefore, this study suggests a new mechanism by which Plk1 regulates MCAK: by regulating its degradation and hence controlling its turnover in mitosis.
Global Governance ist in: Klingt neuartig, global ist eigentlich auch alles und wer will sich schon vorwerfen lassen, noch im „methodologischen Nationalismus“ verhaftet zu sein? Dies ist überspitzt formuliert, doch wie sich zeigte nicht ganz unberechtigt. Nichtsdestotrotz waren die OrganisatorInnen des ersten Weltkongresses der International Association of Political Science Students (IAPSS) wohl selbst überrascht als zur Tagung mit dem Titel „The Limits of Global Governance“ rund 200 Anmeldungen von Studierenden kamen. In Thessaloniki, der europäischen Jugendhauptstadt 2014, wurde vier Tage über Global Governance debattiert und in den insgesamt sieben studentischen Panels, zahlreichen Vorträgen von etablierten WissenschaftlerInnen und weiteren Veranstaltungen zeigte sich vor allem die Vielfalt und Breite des Themas. Wir werden daher im Folgenden Schlaglichter auf interessante Veranstaltungen und Inhalte werfen und abschließend ein kritisch-konstruktives Fazit der Tagung ziehen.
MiR144/451 expression is repressed by RUNX1 during megakaryopoiesis and disturbed by RUNX1/ETO

Abstract: A network of lineage-specific transcription factors and microRNAs tightly regulates differentiation of hematopoietic stem cells along the distinct lineages. Deregulation of this regulatory network contributes to impaired lineage fidelity and leukemogenesis. We found that the hematopoietic master regulator RUNX1 controls the expression of certain microRNAs, of importance during erythroid/megakaryocytic differentiation. In particular, we show that the erythorid miR144/451 cluster is epigenetically repressed by RUNX1 during megakaryopoiesis. Furthermore, the leukemogenic RUNX1/ETO fusion protein transcriptionally represses the miR144/451 pre-microRNA. Thus RUNX1/ETO contributes to increased expression of miR451 target genes and interferes with normal gene expression during differentiation. Furthermore, we observed that inhibition of RUNX1/ETO in Kasumi1 cells and in RUNX1/ETO positive primary acute myeloid leukemia patient samples leads to up-regulation of miR144/451. RUNX1 thus emerges as a key regulator of a microRNA network, driving differentiation at the megakaryocytic/erythroid branching point. The network is disturbed by the leukemogenic RUNX1/ETO fusion product.
Author Summary: The regulatory network between transcription factors, epigenetic cofactors and microRNAs is decisive for normal hematopoiesis. The transcription factor RUNX1 is important for the establishment of a megakaryocytic gene expression program and the concomitant repression of erythroid genes during megakaryocytic differentiation. Gene regulation by RUNX1 is frequently disturbed by mutations and chromosomal translocations, such as the t(8;21) translocation, which gives rise to the leukemogenic RUNX1/ETO fusion protein. We found that RUNX1 regulates microRNAs, which are of importance at the megakaryocytic/erythroid branching point. Specifically, RUNX1 down-regulates expression of the microRNA cluster miR144/451 during megakaryocytic differentiation by changing the epigenetic histone modification pattern at the locus. We could show that miR451, one of the micorRNAs of the miR144/451 cluster, supports erythroid differentiation. We found that expression of miR451 is repressed by the RUNX1/ETO fusion protein, resulting in up regulation of miR451 target genes. Our data support the notion that RUNX1 suppresses the erythroid gene expression program including the erythroid microRNA miR451 and that the RUNX1/ETO fusion protein interferes with normal gene regulation by RUNX1.
Wasserbedarfsprognosen sind für Wasserversorger eine wichtige Entscheidungsgrundlage für zukünftige Maßnahmen in der wirtschaftlichen und technischen Betriebsführung sowie beim Ressourcenmanagement. In den letzten zwei Jahrzehnten sanken in Deutschland die spezifischen Wasserbedarfe aufgrund von Technik- und Verhaltensinnovationen. Für Regionen mit Wirtschafts- und Bevölkerungswachstum ist aber das Zusammenspiel dieser für den zukünftigen Bedarf konträren Entwicklungen von besonderem Interesse. Auch die Metropolregion Hamburg ist von diesen Entwicklungen betroffen.
Im Auftrag des Wasserversorgers HAMBURG WASSER aktualisierte das ISOE (Forschungsschwerpunkt Wasserressourcen und Landnutzung) in Kooperation mit dem ifo Institut München seine mittel- und langfristige Wasserbedarfsprognose aus dem Jahr 2007 für das Versorgungsgebiet des Auftraggebers. In einem innovativen Konzept wurden dafür Forschungsmethoden aus Natur-, Wirtschafts-, Planungs- und Sozialwissenschaften kombiniert. Mit dem gewählten transdisziplinären Forschungsmodus war das Projekt darauf angelegt, im laufenden Forschungsprozess gemeinsam mit den wissenschaftlichen und außerwissenschaftlichen Projektpartnern das Prognosekonzept weiterzuentwickeln. Der vorliegende Studientext basiert auf dem Projektbericht an HAMBURG WASSER und fasst Prognosekonzept, Modellentwicklung, Prognoseergebnissen und Schlussfolgerungen zusammen.
This assessment concept paper provides a methodological approach for the formative assessment and summative assessment of GIZ’s International Water Stewardship Programme (IWaSP) and its component partnerships. IWaSP promotes partnerships between the private sector (corporations and SMEs), the public sector and the society to tackle shared water risks and to manage water equitably to meet competing demands. This evaluative assessment concept describes the generic approach of the assessment, the cycle for the assessment of partnerships, the country coordination and the programme.
The overall goal of the assessment is to provide evidence for taxpayers in the donor countries and for citizens in the partnership countries. It also aims to examine the relevance of the programme’s approach, its underlying assumptions, and the heterogeneity of stakeholders and their specific interests. Since the assessment is also formative feedback to GIZ and IWaSP stakeholders, it aims to guide the future implementation of the partnerships and the programme.
The assessment is guided by several generic principles: assessing for learning (formative assessment); assessment of learning (summative assessment); iteration; structuring complex problems; unblocking results; and conformity with other assessment criteria set out by the OECD the Development Assistance Committee (DAC) and GIZ’s Capacity Works success factors (GTZ 2010).
These generic criteria are adapted to the three levels of the IWaSP structure. First, the assessment cycle for partnerships includes the validation of stakeholders (mapping), the analysis of secondary literature, face-to-face interviews and a process for feeding back the findings. Generic tools are provided to guide the assessment, such as a list of key documents and an interview guide. Partnerships will undergo a baseline, interim assessment and final assessment. As progress varies across individual IWaSP partnerships, the steps taken by each partnership to assess shared water risks, prioritise and agree interventions, are expected to differ slightly. In response to these differences the sequencing and content of the assessment may need to be adapted for the different partnerships.
Second, the country-level assessment considers issues such as the coordination of partnerships within a country, scoping strategies, and interaction between partnership and the programme. Information gathered during the partnership assessment feeds into the country-level assessment.
Third, the assessment cycle for the programme involves a document and monitoring plan analysis, reflection on the different perspectives of the programme staff, country staff and external stakeholders.
The final section is concerned with reporting. Several annexes are provided relating to the organisation and preparation of the assessment, including question guidelines and analysis procedures.
