Chronische Herzinsuffizienz ist eine der führenden Todesursachen im Rahmen kardiovaskulärer Erkrankungen und ist mit einer hohen Anzahl an Komorbiditäten assoziiert. Unter anderem führt Herzinsuffizienz zu Veränderungen des Immunsystems, welche denen der Immunoseneszenz ähneln. Als einflussreiche Modulatoren ganzer molekularbiologischer Regelkreise sind microRNAs in den letzten Jahren zunehmend in den Fokus gerückt. Diese nicht-kodierenden, kurzen RNA-Einzelstränge können die Genexpression von vielen Zielgenen durch spezifisches Binden der jeweiligen mRNA Transkripte kontrollieren. Aufgrund zum Teil gewebe-, zelltyp- und prozess-spezifischer Expression können microRNAs auch als mögliche Biomarker für spezifische klinische Fragestellungen dienen.
Im Rahmen der vorliegenden Arbeit wurde die Expression von immunmodulatorischen microRNAs im peripheren Blut (PB) von jungen und gealterten gesunden Probanden (y/h bzw. o/h) sowie Patienten mit chronischer Herzinsuffizienz (CHF) untersucht. Dabei wurde ein Bezug auf Immunoseneszenz bzw. die Auswirkung von CHF auf das Immunsystem hergestellt. Im Rahmen dessen wurden Leukozyten-, insbesondere Lymphozyten-Subpopulationen analysiert.
Hierzu wurden Probanden in die drei folgenden Gruppen eingeschlossen: Patienten mit CHF (n=18, durchschnittliches Alter 64 Jahre), alters-korrelierte gesunde Probanden (n=13, durchschnittliches Alter 64 Jahre) sowie junge gesunde Probanden (n=30, durchschnittliches Alter 25 Jahre). Neben der Erhebung der klinischen Daten wurde peripheres Blut zur Bestimmung der microRNA-Expressionslevels sowie für durchflusszytometrische Analysen der Leukozytenpopulationen gewonnen.
In den Expressionsanalysen konnte eine alters- und herzinsuffizienz-abhängige Dysregulation einzelner microRNAs beobachtet werden. Insbesondere Mitglieder der miR-181-Familie, spezifisch miR-181a und miR-181c, waren im Alter niedriger exprimiert, zudem war die Expression von miR-181c bei Vorliegen einer CHF deutlicher reduziert. Des Weiteren zeigte sich eine altersabhängige erhöhte Expression von miR-34a, wobei das Vorliegen von CHF keine Auswirkung auf die Expression zeigte. Bei den microRNAs miR-146a und miR-223 konnte keine signifikante alters- oder CHF-abhängige Regulation nachgewiesen werden. Lediglich zeigte bei der miR-155 zeigte sich eine signifikante Reduktion bei Vorliegen einer CHF im Vergleich o/h Probanden.
In Hinblick auf die Leukozytenpopulationen im peripheren Blut wiesen Patienten mit CHF höhere Zahlen an Leukozyten auf, alle miR-181 Mitglieder zeigten hierbei eine inverse Korrelation. Dagegen stellte sich in Hinblick auf die Zusammensetzung der Leukozyten-Subpopulationen eine Reduktion der Lymphozytenfraktion im Alter dar, besonders bei Patienten mit CHF. Insbesondere zeigte sich eine altersabhängige Abnahme der B-Lymphozytenpopulation, wobei auch hier das Vorliegen einer CHF diesen Effekt verstärkte. Die Expression miR-181a und miR-181c sowie miR-146a und miR-223 korrelierte positiv mit dem Anteil der B-Lymphozyten. Innerhalb der B-Zellen zeigte sich eine alters- und CHF-abhängige Reduktion der naïven B-Zellen, welche positiv mit der Expression von miR-181c, miR-146a und miR-223 korrelierte. Die beobachteten Veränderungen der B-Zell-Subpopulationen zeigten sich insbesondere bei CHF Patienten mit ischämischer Ursache im Vergleich zur dilatativen Kardiomyopathie.
Bei den untersuchten T-Lymphozyten-Subpopulationen zeigte sich eine altersabhängiger Abfall bei den zytotoxischen T-Zellen. Das Vorliegen einer CHF verstärkte diesen Effekt. Die beobachteten Veränderungen der T-Zell-Subpopulation korrelierten nicht mit der Expression der untersuchten microRNAs.
Im Gegensatz zu den lymphoiden Subpopulationen zeigte sich ein Anstieg der neutrophilen Granulozyten und der Monozyten im Alter, es stellte sich jeweils eine negative Korrelation mit der Expression von miR-181 Transkripten sowie miR-155, miR-146a und miR-223 dar.
Zusammenfassend zeigten sich signifikant erniedrigte Expressionslevels von miR-181c im Alter, was mit Immunoseneszenz-bedingten Veränderungen des peripheren Bluts einherging. Diese Veränderungen zeigten sich zusätzlich verstärkt bei Patienten mit CHF. Zukünftig könnten miR-181c Expressionslevel in peripherem Blut als Biomarker für die Immunfunktionen bei CHF Patienten dienen und in Hinblick auf eine mögliche prospektive Information evaluiert werden.
Reggie-1 (flotillin-2) and reggie-2 (flotillin-1) are membrane microdomain proteins which are associated with the membrane by means of acylation. They influence different cellular signaling processes, such as neuronal, T-cell and insulin signaling. Upon stimulation of the EGF receptor, reggie-1 becomes phosphorylated and undergoes tyrosine 163 dependent translocation from the plasma membrane to endosomal compartments. In addition, reggie-1 was shown to influence actindependent processes. Reggie-2 has been demonstrated to affect caveolin- and clathrin-independent endocytosis. Both proteins form homo- and hetero-oligomers, but the function of these oligomers has remained elusive. Moreover, it has not been clarified if functions of reggie-1 are also influenced by reggie-2 and vice versa. The first aim of the study was to further investigate the interplay and the heterooligomerization of reggie proteins and their functional effects. Both reggie proteins were individually depleted by means of siRNA. In different siRNA systems and various cell lines, reggie-1 depleted cells showed reduced protein amounts of reggie-1 and reggie-2, but reggie-2 knock down cells still expressed reggie-1 protein. The decrease of reggie-2 in reggie-1 depleted cells was only detected at protein but not at mRNA level. Furthermore, reggie-2 expression could be rescued by expression of siRNA resistant wild type reggie-1-EGFP constructs, but not by the soluble myristoylation mutant G2A. This mutant was also not able to associate with endogenous reggie-1 or reggie-2, which demonstrates that membrane association of reggie-1 is necessary for hetero-oligomerization. In addition, fluorescence microscopy studies and membrane fractionations showed that correct localization of overexpressed reggie-2 was dependent on co-overexpressed reggie-1. Thus, hetero-oligomerization is crucial for membrane association of reggie-2 and for its protein stability or protein expression. Moreover, the binding of reggie-2 to reggie-1 required tyrosine 163 of reggie-1 which was previously shown to be important for endosomal translocation of reggie-1. Since reggie-2 was implicated to function in clathrin- and caveolin-independent endocytosis pathways, the effect of reggie-2 depletion on reggie-1 endocytosis was investigated. Indeed, reggie-1 was dependent on reggie-2 for endosomal localization and EGF-induced endocytosis. By FRET-FLIM analysis it could be shown that reggie heterooligomers are dynamic in size or conformation upon EGF stimulation. Thus, it can be concluded that reggie proteins are interdependent in different aspects, such as protein stability or expression, membrane association and subcellular localization. In addition, these results demonstrate that the hetero-oligomers are dynamic and reggie proteins influence each other in terms of function. A further aim was the characterization of reggie-1 and reggie-2 function in actindependent processes, where so far only reggie-1 was known to play a role. Depletion of either of the proteins reduced cell migration, cell spreading and the number of focal adhesions in steady state cells. Thus, also reggie-2 affects actin-dependent processes. Further investigation of the focal adhesions during cell spreading revealed that depletion of reggie-1 displayed different effects as compared to reggie-2 knock down. Reggie-1 depleted cells had elongated cell-matrix-adhesions and showed reduced activation of FAK and ERK2. On the other hand, depletion of reggie-2 resulted in a restricted localization of focal adhesion at the periphery of the cell and decreased ERK2 phosphorylation, but it did not affect FAK autophosphorylation. Hence, reggie proteins influence the regulation of cell-matrix-adhesions differently. A link between reggie proteins and focal adhesions is the actin cross-linking protein -actinin. The interaction of -actinin with reggie-1 could be verified by means of co-immunoprecipitations and FRET-FLIM analysis. Reggie-1 binds -actinin especially in membrane ruffles and in other locations where actin remodeling takes place. Moreover, -actinin showed a different localization pattern during cell spreading in reggie-1 depleted cells, as compared to the control cells. These results provide further insights into the function of both reggie proteins. Their interplay and hetero-oligomerization was shown to be crucial for their role in endocytosis. In addition, both reggie proteins influence actin-dependent processes and differentially affect focal adhesion regulation.
This work is about resumptive and non-resumptive relative clauses (RCs) in the three big Ibero-Romance languages: Spanish, Portuguese, and Catalan. In (1), the examined structures are exemplified for Spanish: (1a.) No conozco el hombre que viste _ ayer. “I don’t know the man that you saw yesterday.” (1b.) Es este el hombre que le enviaron el libro. “This is the man to whom they sent the book.” (1c.) Es este el hombre a quien le enviaron el libro. “This is the man to whom they sent the book.”
(1a.) displays a non-resumptive, or canonical, RC, which is characterized by the canonical use of a relativizing operator and a gap in the subordinate’s object position, a piece of evidence which has induced most of the generative literature to assume wh-movement of the relative operator in the sense of Chomsky (1977). The last two decades, however, have seen a big debate regarding the exact derivational analysis, starting with Kayne’s (1994) antisymmetry theory and the following focus on reconstruction and anti-reconstruction effects in RCs. This search for the correct starting site of the RC’s head noun has dismissed the original Head External Analysis (HEA) (Chomsky 1965, 1977) and led to the development of a Head Raising Analysis (RA) (Kayne 1994, Bianchi 1999, a.o.) and a Matching Analysis (MA) (Munn 1994, Sauerland 1998, a.o.). The discussion in this work argues that the data on reconstruction and anti-reconstruction effects are not sufficiently clear and reliable in order to adopt one of the head-internal analyses, i.e. a HEA or a MA. Instead, the work follows a variant of the HEA proposed for Portuguese by Rinke & Aßmann (2017), which adheres to standard assumptions about Romance syntax, and avoids the empirical problems that the other proposals have to face. Arguing that the HEA holds for all Ibero-Romance languages, this work also takes a stance in the debate around the categorical status of the relativizing element que and argues that it is always a D-element, and never of category C, i.e. there is no such thing as a relativizing complementizer (cf. also Kayne 2010, Kato & Nunes 2008, Poletto & Sanfelici 2018).
The work argues that wh-movement in a HEA fashion is the correct analysis also for resumptive relative clauses as in (1b., c.), which crucially lack a gap in argument position but show a resumptive pronominal element instead. Furthermore, it takes advantage of the fact that the choice of such genetically closely related languages like Spanish, Portuguese, and Catalan enables research to address the phenomenon under consideration from a microcomparative perspective, which is “the closest we can come … to a controlled experiment in comparative syntax” (Kayne 2005: 281-282). The descriptive literature suggests that, at least for Spanish and Catalan, there are two types of a resumptive RC structure available: a simple resumption as in (1b.), including mere que, and a complex resumption structure which displays a more complex relativizer like a quien in combination with a resumptive pronoun (1c.). However, a corpus study carried out for this work reveals that speakers of the three languages behave alike insofar as the only resumptive RC used in spontaneous speech is a simple-resumption structure, while complex resumption never occurs. Additionally, a multivariate analysis shows that in all three languages, grammatical case is the most important factor when it comes to the possibility of a resumptive structure in RCs: with a dative argument, simple resumption is obligatory, while for accusative and nominative arguments, resumption is optional. The discussion concludes that simple and complex resumption constitute different phenomena also on a structural level: the latter one is argued to be a subcase of clitic doubling, and therefore, receives an analysis along the lines of Pineda (2016), who argues against a dative alternation in Romance languages and locates the (non-)realisation of the dative clitic in a transitive clitic-doubling structure outside of syntax, it being a case of silent variation along the lines of Sigurðsson (2004) and Kayne (2005). From this perspective, it follows naturally that in Portuguese, complex resumption structures are ungrammatical. Simple resumption, on the other hand, which is a possible structure in all three languages, is argued to represent the phonological counterpart of “scattered deletion”, i.e. the preferred interpretation for an A’-chain according to Chomsky (2003): in the operator position SpecCP, every feature except for the operator feature is deleted, resulting in the phonological outcome que, while in the variable position, everything but the operator is interpreted, resulting in a pronominal element according to the argument’s phi-features.
This work takes a stance in the latest topics on generative analyses for relative clauses. Using not only theoretical considerations but conclusions drawn from empirical data on three languages, it offers a new perspective on pending questions and proposes to take a fresh look on supposedly outdated analyses.
Alzheimer’s disease (AD) is the most common neurodegenerative disorder world wide, causing presenile dementia and death of millions of people. During AD damage and massive loss of brain cells occur. Alzheimer’s disease is genetically heterogeneous and may therefore represent a common phenotype that results from various genetic and environmental influences and risk factors. In approximately 10% of patients, changes of the genetic information were detected (gene mutations). In these cases, Alzheimer’s disease is inherited as an autosomal dominant trait (familial Alzheimer’s disease, FAD). In rare cases of familial Alzheimer’s disease (about 1-3%), mutations have been detected in genes on chromosomes 14 and 1 (encoding for Presenilin 1 and 2, respectively), and on chromosome 21 encoding for the amyloid precursor protein (APP), which is responsible for the release of the cell-damaging protein amyloid-beta (ß-amyloid, Aß). Familial forms of early-onset Alzheimer’s disease are rare; however, their importance extends far beyond their frequency, because they allow to identify some of the critical pathogenetic pathways of the disease. All familial Alzheimer mutations share a common feature: they lead to an enhanced production of the Aß, which is the major constituent of senile plaques in brains of AD patients. New data indicates that Aß promotes neuronal degeneration. Therefore, one aim of these thesis was to elucidate the neurotoxic biochemical pathways induced by Aß, investigating the effect of the FAD Swedish APP double mutation (APPsw) on oxidative stress-induced cell death mechanisms. This mutation results in a three- to sixfold increased Aß production compared to wild-type APP (APPwt). As cell models, the neuronal PC12 (rat pheochromocytoma) and the HEK (human embryonic kidney 293) cell lines were used, which have been transfected with human wiltyp APP or human APP containing the Swedish double mutation. The used cell models offer two important advantages. First, compared to experiments using high concentrations of Aß at micromolar levels applied extracellularly to cells, PC12 APPsw cells secret low Aß levels similar to the situation in FAD brains. Thus, this cell model represents a very suitable approach to elucidate the AD-specific cell death pathways mimicking physiological conditions. Second, these two cell lines (PC12 and HEK APPwt and APPsw) with different production levels of Aß may additionally allow to study dose-dependent effects of Aß. The here obtained results provide evidence for the enhanced cell vulnerability caused by the Swedish APP mutation and elucidate the cell death mechanism probably initiated by intracellulary produced Aß. Here it seems likely that increased production of Aß at physiological levels primes APPsw PC12 cells to undergo cell death only after additional stress, while chronic high levels in HEK cells already lead to enhanced basal apoptotic levels. Crucial effects of the Swedish APP mutation include the impairments of cellular energy metabolism affecting mitochondrial membrane potential and ATP levels as well as the additional activation of caspase 2, caspase 8 and JNK in response to oxidative stress. Thereby ,the following model can be proposed: PC12 cells harboring the Swedish APP mutation have a reduced energy metabolism compared to APPwt or control cells. However, this effect does not leads to enhanced basal apoptotic levels of cultured cells. An exposure of PC12 cells to oxidative stress leads to mitochondrial dysfunction, e.g., decrease in mitochondrial membrane potential and depletion in ATP. The consequence is the activation of the intrinsic apoptotic pathway releasing cytochrome c and Smac resulting in the activation of caspase 9. This effect is amplified by the overexpression of APP, since both APPsw and APPwt PC12 cells show enhanced cytochrome c and Smac release as well as enhanced caspase 9 activity as vector transfected control. In APPsw PC12 cells a parallel pathway is additionally emphased. Due to reduced ATP levels or enhanced Aß production JNK is activated. Furthermore, the extrinsic apoptotic pathway is enhanced, since caspase 8 and caspase 2 activation was clearly enhanced by the Swedish APP mutation. Both pathways may then converge by activating the effector enzyme, caspase 3, and the execution of cell death. In addition, caspase independent effects also needs to be considered. One possibility could be the implication of AIF since AIF expression was found to be induced by the Swedish APP mutation. In APPsw HEK cells high chronic Aß levels leads to enhanced apoptotic levels, reduce mitochondrial membrane potential and ATP levels even under basal conditions. Summarizing, a hypothetical sequence of events is proposed linking FAD, Aß production, JNK-activation, mitochondrial dysfunction with caspase pathway and neuronal loss for our cell model. The brain has a high metabolic rate and is exposured to gradually rising levels of oxidative stress during life. In Swedish FAD patients the levels of oxidative stress are increased in the temporal inferior cortex. This study using a cell model mimicking the in vivo situation in AD brains indicates that probably both, increased Aß production and the gradual rise of oxidative stress throughout life converge at a final common pathway of an increased vulnerability of neurons to apoptotic cell death from FAD patients. Presenilin (PS) 1 is an aspartyl protease, involved in the gamma-secretase mediated proteolysis of Amyloid-ß-protein (Aß), the major constituent of senile plaques in brains of Alzheimer’s disease (AD) patients. Recent studies have suggested an additional role for presenilin proteins in apoptotic cell death observed in AD. Since PS 1 is proteolytic cleaved by caspase 3, it has been prosposed that the resulting C-terminal fragment of PS1 (PSCas) could play a role in signal transduction during apoptosis. Moreover, it was shown that mutant presenilins causing early-onset of familial Alzheimer's disease (FAD) may render cells vulnerable to apoptosis. The mechanism by which PS1 regulates apoptotic cell death is yet not understood. Therefore one aim of our present study was to clarify the involvement of PS1 in the proteolytic cascade of apoptosis and if the cleavage of PS1 by caspase 3 has an regulatory function. Here it is demonstrated that both, PS1 and PS1Cas lead to a reduced vulnerability of PC12 and Jurkat cells to different apoptotic stimuli. However a mutation at the caspase 3 recognition site (D345A/ PSmut), which inhibits cleavage of PS1 by caspase 3, show no differences in the effect of PS1 or PSCas towards apoptotic stimuli. This suggest that proteolysis of PS1 by caspase 3 is not a determinant, but only a secondary effect during apoptosis. Since several FAD mutation distributed through the whole PS1 gene lead to enhanced apoptosis, an abolishment of the antiapoptotic effect of PS1 might contribute to the massive neurodegeneration in early age of FAD patients. Here, the regulate properties of PS1 in apoptosis may not be through an caspase 3 dependent cleavage and generation of PSCas, but rather through interaction of PS1 with other proteins involved in apoptosis.
Diese Arbeit etabliert eine nicht-invasive, volloptische Methode zur in-vivo Beobachtung des Membranpotentials in erregbaren Zellen des Fadenwurms C. elegans, die als Ersatz oder komplementär zu invasiven, elektrophysiologischen Methoden verwendet werden kann.
Zahnfarbene, ästhetisch unauffällige Brackets bestehen meist aus Keramik oder Kunststoff. Besonders Keramikbrackets weisen durch den starken Haftverbund zum Zahn ein gewisses Risiko der Gefährdung von Zahnhartsubstanz beim Debonding auf. Klinisch sollte eine Haftkraft von 5-10 MPa erreicht werden, damit die einwirkenden therapeutischen Kräfte nicht zum Verlust des Brackets führen. Ziel der Studie war es, ein neuartiges selbstligierendes Bracket (Opal/Ultradent) aus glasfaserverstärktem Kunststoff auf die Haftfestigkeit und das Risikopotenzial beim Debonding zu testen und mit Keramik-Brackets und einem weit verbreiteten Standard-Metall-Bracket zu vergleichen. Es wurden neben dem Opal-Bracket (Ultradent) vier Keramik-Brackets (Clarity/ 3M, Fascination2/ Dentaurum, Aspire/ Forestadent, Inspire Ice/ Ormco) und ein Metall-Bracket (Victory/ 3M) mit dem vom Hersteller empfohlenen Adhäsivsystem getestet. Die zur Untersuchung der Haftfestigkeit verwendeten 120 humanen Weisheitszähne wurden für 7 Tage in einer 0.5 % Chloramin T-Lösung und anschließend in Wasser gelagert und danach nochmals mikroskopisch auf Unversehrtheit des Zahnschmelzes überprüft. Mit einer Messschablone wurde der Krümmungsradius der bukkalen Flächen überprüft. Der Krümmungsradius der Zahnoberfläche durfte nicht kleiner als 12,5mm sein. Nach gründlicher Reinigung mit Bimsmehl wurden die Weisheitszähne auf sechs Gruppen mit jeweils 20 Brackets aufgeteilt. Die Brackets wurden mit dem vom Hersteller empfohlenem Adhäsiv- Primer-System nach Herstellerangaben auf die Schmelzoberfläche geklebt. Alle Prüfkörper wurden mit einer Einbetthilfe in Palapress Vario (Heraeus Kulzer) eingebettet und anschließend in einer selbst hergestellten Justierhilfe einheitlich ausgerichtet. Nach einer Lagerungszeit von 24 Stunden in Wasser bei 37° C erfolgte die Abscherung der Prüfkörper nacheinander im rechten Winkel zur Bracketbasis mit der Zwick-Universalprüfmaschine (Ulm, Germany). Die Vorschubgeschwindigkeit betrug 1mm/min. Der Weg bis zum Abscheren und die dazu benötigte Kraft eines jeden Prüfkörpers wurden digital aufgezeichnet. Anhand der Fläche der Bracketbasis, der Abscherkraft und dem dazu benötigten Weg konnten die Haftfestigkeiten in MPa umgerechnet und miteinander verglichen werden. Der Gruppenvergleich erfolgte mit dem ANOVA und Scheffe-Test. Die Zahnoberflächen wurden zusätzlich mikroskopisch auf Schmelzdefekte überprüft. Das Metall-Bracket, das als Vergleichs- und Normierungsgröße diente, zeigte eine mittlere Haftfestigkeit von 10,2 MPa. Das Opal-Bracket wies mit 4,17 MPa die signifikant niedrigste durchschnittliche Haftkraft auf. Es erreichte nicht die klinisch ausreichende Haftkraft von 6 MPa. Die Keramik-Brackets erzielten mit 14.7 MPa (Clarity), 15,3 MPa (Aspire) und 14.7 MPa (Inspire Ice) ähnliche Haftwerte, ohne zu Hartsubstanzdefekten zu führen. Bei Fascination2 mit einer durchschnittlichen Haftkraft von 19,7 MPa konnten nach dem Debonding bei ca. 1/3 der Proben Schmelzausrisse nachgewiesen werden. Fascination2 zeigte im Vergleich zur Kontrollgruppe signifikant höhere Haftwerte (19,7 MPa). Die Haftfestigkeit des Opal-Brackets scheint im Vergleich zu den in der Literatur geforderten Angaben für die sichere klinische Anwendung zu gering. Die Haftkraft von Fascination2 (Dentaurum) ist durch die zeitweise auftretenden Schmelzausrisse für ein sicheres Debonding als zu hoch zu bewerten. Die anderen Keramik-Brackets zeigen ein gutes Verhältnis zwischen sicherer Entfernbarkeit und ausreichendem Haftverbund und bieten eine Alternative zu den Metallbrackets.
Einleitung: Die akute Tonsillitis gehört zu den Infektionen der oberen Atemwege und ist eine sehr häufige Erkrankung in einer Kinder- und Jugendarztpraxis. Ziel unserer Untersuchung war es, das virale und bakterielle Erregerspektrum der akuten Tonsillitis, ihre saisonale Verteilung, ihre Altersverteilung, die klinische Symptomatik und den Einfluss einer Rauchexposition zu untersuchen. Gleichzeitig sollte erneut die Sensitivität und Spezifität des angewandten StrepA ST überprüft werden.
Methoden: In drei Kinder- und Jugendarztpraxen im Rhein-Main Gebiet wurden zwischen April 2009 und Mai 2010 insgesamt 1720 Patienten mit akuten Halsschmerzen untersucht. Mit einem Anamnesebogen wurden Alter, klinische Symptomatik und die Rauchexposition erfasst. Bei allen Patienten wurde ein StrepA ST durchgeführt. In einer Praxis (Praxis 1) wurden bei 306 Patienten zusätzlich ein Abstrich für eine bakterielle Kultur und für eine Multiplex PCR auf Viren durchgeführt.
Resultate: In 84% der Fälle tritt die GAS Tonsillitis im Alter zwischen 2 und 12 Jahren auf. Es konnte keine saisonale Häufung der GAS nachgewiesen werden. Mit 64 (42%) StrepA ST positiven Patienten von 152 Raucher-Familien und 74 (37%) von 200 Nichtraucher-Raucher-Familien zeigt sich kein signifikant erhöhtes Risiko an einer GAS Tonsillitis zu erkranken, wenn mindestens ein Elternteil raucht. Bei 306 Rachenabstrichen konnten 145 (47,5%) mal Streptokokken nachgewiesen werden. Davon waren mit 133 vorwiegend GAS (92%). Die anderen Streptokokken der Gruppen C (4,8%), G (2,1%) und B (1,4%) kommen deutlich seltener vor und spielen eine untergeordnete Rolle. Die Sensitivität und Spezifität des StrepA ST war mit 89,9% und 94,1% ausgezeichnet. Bei 306 Tonsillitiden gelang bei 110 Patienten (35,8%) ein Virusnachweis. Wie erwartet fanden sich doppelt so viele Virusnachweise (46%) bei Patienten ohne GAS Nachweis als Ko-Infektionen bei einer GAS (24%). Der Anteil der Entero-/Rhinoviren unter den nachgewiesenen Viren war mit 54% am höchsten. Adenoviren waren mit 15% und Influenza- 9% die nächst häufigen Viren, Para- und Coronaviren bildeten kleinere Gruppen. Während Entero-/Rhinoviren ganzjährig vorkommen sind Influenzaviren eher in der kalten Jahreszeit für akute Tonsillitiden verantwortlich. Die Rolle der Ko-Infektion in der Entstehung und im Verlauf der akuten Tonsillitis muss in weiteren Untersuchungen erforscht werden.
Die vorliegende Arbeit beschäftigt sich mit unterschiedlichen Aspekten der Faltung von Tendamistat, einem aus 74 Aminosäuren bestehenden a-Amylase lnhibitor aus Streptomyces tendae. Bei der oxidativen Rückfaltung des Tendamistats konnten bisher drei kinetische Phasen (t1, t2 und t3) identifiziert werden. Die mittlere (t2) bzw. langsame (ti) kinetische Phase, resultiert als Folge einer cis/trans lsomerisierung um eine der drei Xaa-Pro Bindungen im Tendamistat. Wie im ersten Teil dieser Arbeit gezeigt werden konnte, entsteht die prolinabhängige lsomerisierungsreaktion während der Rückfaltung (t3) als Folge einer trans -> cis Umlagerung um die A1a49-Pro50 Amidbindung nach Entfaltung des Proteins. Der Nachweis einer solchen lsomerisierung gelang auf rekombinantem Weg, durch gezielten Austausch der einzelnen Proline. Im Gegensatz dazu resultiert die mittlere kinetischen Phase (t2) als Folge einer Prolin-unabhängigen cis/trans-lsomerisierungsreaktion um mindestens eine vorhandenen nicht-Prolinbindungen im Protein. Diese Schlußfolgerung konnte durch Vergleich mit zahlreichen Literaturdaten und Experimenten weiter gestützt werden. Tendamistat ist damit das erste Protein in der Literatur, bei dem es gelang, eine solche Prolin-unabhängige lsomerisierungsreaktion experimentell nachzuweisen. Wie mit Hilfe von weiteren Substitutionsexperimenten im zweiten Teil der Arbeit gezeigt werden konnte, tragen hydrophobe Oberflächencluster entscheidend zur Stabilisierung des lnhibitors bei. Die dabei gemessenen Stabilisierungsbeiträge stehen in einem linearen Verhältnis zum Seitenkettenvolumen der eingeführten Aminosäurereste (Alanin << Valin << Isoleucin). Hierdurch ließ sich zeigen, daß der stabilisierende Einfluß solcher Cluster im Wesentlichen auf das Vorhandensein von hydrophoben Wechselwirkungen zurückzuführen ist. Für die Faltung von Proteinen, die wie das Tendamistat ausschließlich aus ß-Faltblättern bestehen, spielt die Bildung von ß-Hairpinstrukturen eine entscheidende Rolle Die in dieser Arbeit untersuchten ß-Hairpinmutanten L14A bzw. N25A zeigen im Vergleich zum Wildtyp eine extreme Destabilisierung der ß-Faltblattstruktur. Durch Vergleich der Aktivierungsenergien (DeltadeltaGD-TS) mit den freien Enthalpien (DeltadeltaG0N-D) konnte die strukturelle Entwicklung des ß-Hairpins im Übergangszustand an den beiden Positionen 14 bzw. 25 mitverfolgt werden (~-Wert Analyse). Entgegen den Erwartungen zeigen diese Untersuchungen, daß beide Positionen im Übergangszustand noch weitestgehend solvatisiert vorliegen. Damit scheint die nativ-ähnliche Zusammenlagerung des ersten ß-Hairpins erst nach Erreichen des Übergangszustands stattzufinden. Alle in dieser Arbeit vorgelegten Daten über die Faltung des alpha-Amylase lnhibitors sind konsistent mit dem Nukleations-Kondensations Modell. Analog der Tröpfchenbildung bei unterkühlten Gasen kommt es nach dem kollabieren der Proteinkette als Reaktion auf die veränderten Lösungsmittelbedingungen zur intramolekularen Suche nach Nukleationsstellen. Der Übergangszustand wird erreicht, wenn sich eine kritische Zahl stabilisierender Kontakte (unspezifische Nukleation) ausbilden konnte. Ob an einem solchen Nukleationskern stets bestimmte Seitenketten beteiligt sind (spezifische Nukleation), bleibt an dieser Stelle noch offen.
Nematophilic bacteria as a source of novel macrocyclised antimicrobial non-ribosomal peptides

A solution to ineffective clinical antimicrobials is the discovery of new ones from under-explored sources such as macrocyclic non-ribosomal peptides (NRP) from nematophilic bacteria. In this dissertation an antimicrobial discovery process –from soil sample to inhibitory peptide– is demonstrated through investigations on six nematophilic bacteria: Xenorhabdus griffiniae XN45, X. griffiniae VH1, Xenorhabdus sp. nov. BG5, Xenorhabdus sp. nov. BMMCB, X. ishibashii and Photorhabdus temperata. To demonstrate the first step of bacterium isolation and species delineation, endosymbionts were isolated from Steinernema sp. strains BG5 and VH1 that were isolated directly from soil samples in Western Kenya. After genome sequencing and assembly of novel Xenorhabdus isolates VH1 and BG5, species delineation was done via three overall genome relatedness indices. VH1 was identified as X. griffiniae VH1, BG5 as Xenorhabdus sp. nov. BG5 and X. griffiniae BMMCB was emended to Xenorhabdus sp. nov. BMMCB. The nematode host of X. griffiniae XN45, Steinernema sp. scarpo was highlighted as a putative novel species. To demonstrate the second step of genome mining and macrocyclic non-ribosomal peptide structure elucidation, chemosynthesis and biosynthesis, the non-ribosomal peptide whose production is encoded by the ishA-B genes in X. ishibashii was investigated. Through a combination of refactoring the ishA-B operon by a promoter exchange mechanism, isotope labelling experiments, high resolution tandem mass spectrometry analysis, bioinformatic protein domain analysis and chemoinformatic comparisons of actual to hypothetical mass spectrometry spectra, the structures of Ishipeptides were elucidated and confirmed by chemical synthesis. Ishipeptide A was a branch cyclic depsidodecapeptide macrocyclised via an ester bond between serine and the terminal glutamate. It chemosynthesis route was via a late stage macrolactamation and linearised Ishipeptide B was synthesised via solid phase iterative synthesis. Ishipeptides were not N-terminally acylated despite being biosynthesised from the IshA protein that had a C-starter domain. It was highlighted that more than restoration of the histidine active site of this domain is required to restore N-terminal acylation activity.
To demonstrate the final step of determination of antimicrobial activity, minimum inhibitory concentrations of Ishipeptides and Photoditritide from Photorhabdus temperata against fungi and bacteria were determined. None were antifungal while only the macrocyclic compounds were inhibitory, with Ishipeptide A inhibitory to Gram-positive bacteria at 37 µM. The cationic Photoditritide, a cyclic hexapeptide macrocyclised via a lactam bond between homoarginine and tryptophan, was 12 times more inhibitory (3.0 µM), even more effective than a current clinical compound, Ampicillin (4.2 µM). For both, macrocyclisation was hypothesised to contribute to antimicrobial activity. Ultimately, this dissertation demonstrated not only nematophilic bacteria as a source of novel macrocyclic antimicrobial non-ribosomal peptides but also a process of antimicrobial discovery–from soil sample to inhibitory peptide– from these useful bacteria genera. This is significant for the fight against antimicrobial resistance.
