First measurements of hadron(h)−Λ azimuthal angular correlations in p−Pb collisions at sNN−−−√ = 5.02 TeV using the ALICE detector at the LHC are presented. These correlations are used to separate the production of associated Λ baryons into three different kinematic regions, namely those produced in the direction of the trigger particle (near-side), those produced in the opposite direction (away-side), and those whose production is uncorrelated with the jet-axis (underlying event). The per-trigger associated Λ yields in these regions are extracted, along with the near- and away-side azimuthal peak widths, and the results are studied as a function of associated particle pT and event multiplicity. Comparisons with the DPMJET event generator and previous measurements of the ϕ(1020) meson are also made. The final results indicate that strangeness production in the highest multiplicity p−Pb collisions is enhanced relative to low multiplicity collisions in the jet-like regions, as well as the underlying event. The production of Λ relative to charged hadrons is also enhanced in the underlying event when compared to the jet-like regions. Additionally, the results hint that strange quark production in the away-side of the jet is modified by soft interactions with the underlying event.
Measurements of (anti)deuteron and (anti)3He production in the rapidity range |y|< 0.5 as a function of the transverse momentum and event multiplicity in Xe−Xe collisions at a center-of-mass energy per nucleon−nucleon pair of sNN−−−√ = 5.44 TeV are presented. The coalescence parameters B2 and B3 are measured as a function of the transverse momentum per nucleon. The ratios between (anti)deuteron and (anti)3He yields and those of (anti)protons and pions are reported as a function of the mean charged-particle multiplicity density, and compared with two implementations of the statistical hadronization model (SHM) and with coalescence predictions. The elliptic flow of (anti)deuterons is measured for the first time in Xe−Xe collisions and shows features similar to those already observed in Pb−Pb collisions, i.e., the mass ordering at low transverse momentum and the meson−baryon grouping at intermediate transverse momentum. The production of nuclei is particularly sensitive to the chemical freeze-out temperature of the system created in the collision, which is extracted from a grand-canonical-ensemble-based thermal fit, performed for the first time including light nuclei along with light-flavor hadrons in Xe−Xe collisions. The extracted chemical freeze-out temperature Tchem = (154.2 ± 1.1) MeV in Xe−Xe collisions is similar to that observed in Pb−Pb collisions and close to the crossover temperature predicted by lattice QCD calculations.
The transverse momentum (pT) differential production cross section of the promptly-produced charm-strange baryon Ξ0c (and its charge conjugate Ξ0c¯¯¯¯¯¯) is measured at midrapidity via its hadronic decay into π+Ξ− in p−Pb collisions at a centre-of-mass energy per nucleon−nucleon collision sNN−−−√ = 5.02 TeV with the ALICE detector at the LHC. The Ξ0c nuclear modification factor (RpPb), calculated from the cross sections in pp and p−Pb collisions, is presented and compared with the RpPb of Λ+c baryons. The ratios between the pT-differential production cross section of Ξ0c baryons and those of D0 mesons and Λ+c baryons are also reported and compared with results at forward and backward rapidity from the LHCb Collaboration. The measurements of the production cross section of prompt Ξ0c baryons are compared with a model based on perturbative QCD calculations of charm-quark production cross sections, which includes only cold nuclear matter effects in p−Pb collisions, and underestimates the measurement by a factor of about 50. This discrepancy is reduced when the data is compared with a model in which hadronisation is implemented via quark coalescence. The pT-integrated cross section of prompt Ξ0c-baryon production at midrapidity extrapolated down to pT = 0 is also reported. These measurements offer insights and constraints for theoretical calculations of the hadronisation process. Additionally, they provide inputs for the calculation of the charm production cross section in p−Pb collisions at midrapidity.
Investigating strangeness enhancement with multiplicity in pp collisions using angular correlations

A study of strange hadron production associated with hard scattering processes and with the underlying event is conducted to investigate the origin of the enhanced production of strange hadrons in small collision systems characterised by large charged-particle multiplicities. For this purpose, the production of the single-strange meson K0S and the double-strange baryon Ξ± is measured, in each event, in the azimuthal direction of the highest-pT particle (``trigger" particle), related to hard scattering processes, and in the direction transverse to it in azimuth, associated with the underlying event, in pp collisions at s√=5.02 TeV and s√=13 TeV using the ALICE detector at the LHC. The per-trigger yields of K0S and Ξ± are dominated by the transverse-to-leading production (i.e., in the direction transverse to the trigger particle), whose contribution relative to the toward-leading production is observed to increase with the event charged-particle multiplicity. The transverse-to-leading and the toward-leading Ξ±/K0S yield ratios increase with the multiplicity of charged particles, suggesting that strangeness enhancement with multiplicity is associated with both hard scattering processes and the underlying event. The relative production of Ξ± with respect to K0S is higher in transverse-to-leading processes over the whole multiplicity interval covered by the measurement. The K0S and Ξ± per-trigger yields and yield ratios are compared with predictions of three different phenomenological models, namely PYTHIA 8.2 with the Monash tune, PYTHIA 8.2 with ropes and EPOS LHC. The comparison shows that none of them can quantitatively describe either the transverse-to-leading or the toward-leading yields of K0S and Ξ±.
The first measurement of the impact-parameter dependent angular anisotropy in the decay of coherently photoproduced ρ0 mesons is presented. The ρ0 mesons are reconstructed through their decay into a pion pair. The measured anisotropy corresponds to the amplitude of the cos(2ϕ) modulation, where ϕ is the angle between the two vectors formed by the sum and the difference of the transverse momenta of the pions, respectively. The measurement was performed by the ALICE Collaboration at the LHC using data from ultraperipheral Pb−Pb collisions at a center-of-mass energy of sNN−−−√ = 5.02 TeV per nucleon pair. Different impact-parameter regions are selected by classifying the events in nuclear-breakup classes. The amplitude of the cos(2ϕ) modulation is found to increase by about one order of magnitude from large to small impact parameters. Theoretical calculations, which describe the measurement, explain the cos(2ϕ) anisotropy as the result of a quantum interference effect at the femtometer scale that arises from the ambiguity as to which of the nuclei is the source of the photon in the interaction.
Beyond well-established difficulties with working memory in individuals with attention deficit hyperactivity disorder (ADHD), evidence is emerging that other memory processes may also be affected. We investigated, first, which memory processes show differences in adults and adolescents with ADHD in comparison to control participants, focusing on working and short-term memory, initial learning, interference, delayed and recognition memory. Second, we investigated whether ADHD severity, co-occurring depressive symptoms, IQ and physical fitness are associated with the memory performance in the individuals with ADHD.
We assessed 205 participants with ADHD (mean age 25.8 years, SD 7.99) and 50 control participants (mean age 21.1 years, SD 5.07) on cognitive tasks including the digit span forward (DSF) and backward (DSB), the Rey Auditory Verbal Learning Test (RAVLT), and the vocabulary and matrix reasoning subtests of the Wechsler Abbreviated Scale of Intelligence. Participants with ADHD were additionally assessed on ADHD severity, depression symptoms and cardiorespiratory fitness. A series of regressions were run, with sensitivity analyses performed when variables were skewed.
ADHD-control comparisons were significant for DSF, DSB, delayed and recognition memory, with people with ADHD performing less well than the control participants. The result for recognition memory was no longer significant in sensitivity analysis. Memory performance was not associated with greater ADHD or depression symptoms severity. IQ was positively associated with all memory variables except DSF. Cardiorespiratory fitness was negatively associated with the majority of RAVLT variables.
Individuals with ADHD showed difficulties with working memory, short-term memory and delayed memory, as well as a potential difficulty with recognition memory, despite preserved initial learning.
Targeted protein degradation (TPD) has recently emerged as an exciting new drug modality. However, the strategy of developing small molecule-based protein degraders has evolved over the past two decades and has now established molecular tags that are already in clinical use, as well as chimeric molecules, PROteolysis TArgeting Chimeras (PROTACs), based mainly on ligand systems developed for the two E3 ligases CRBN and VHL. The large size of the human E3 ligase family suggests that PROTACs can be developed by targeting a large diversity of E3 ligases, some of which have restricted expression patterns with the potential to design disease- or tissue-specific degraders. Indeed, many new E3 ligands have been published recently, confirming the druggability of E3 ligases. This review summarises recent data on E3 ligases and highlights the challenges in developing these molecules into efficient PROTACs rivalling the established degrader systems.
he first measurement of 3ΛH and 3Λ¯¯¯¯H¯¯¯¯ differential production with respect to transverse momentum and centrality in Pb−Pb collisions at sNN−−−√=5.02~TeV is presented. The 3ΛH has been reconstructed via its two-charged-body decay channel, i.e., 3ΛH→3He+π−. A Blast-Wave model fit of the pT-differential spectra of all nuclear species measured by the ALICE collaboration suggests that the 3ΛH kinetic freeze-out surface is consistent with that of other nuclei. The ratio between the integrated yields of 3ΛH and 3He is compared to predictions from the statistical hadronisation model and the coalescence model, with the latter being favoured by the presented measurements.
Six C-even states, denoted as X, with quantum numbers JPC=0−+, 1±+, or 2±+, are searched for via the e+e−→γD±sD∗∓s process using (1667.39±8.84) pb−1 of e+e− collision data collected with the BESIII detector operating at the BEPCII storage ring at center-of-mass energy of s√=(4681.92±0.30) MeV. No statistically significant signal is observed in the mass range from 4.08 to 4.32 GeV/c2. The upper limits of σ[e+e−→γX]⋅B[X→D±sD∗∓s] at a 90% confidence level are determined.
Isothermal titration calorimetry (ITC) is a widely used technique for the characterization of protein-protein and protein-ligand interactions. It provides information on the stoichiometry, affinity, and the thermodynamic driving forces of interactions. This chapter exemplifies the use of ITC to investigate interactions between human autophagy modifiers (LC3/GABARAP proteins) and their interaction partners, the LIR motif containing sequences. The purpose of this report is to present a detailed protocol for the production of LC3/GABARAP-interacting LIR peptides using E. coli expression systems. In addition, we outline the design of ITC experiments using the LC3/GABARAP:peptide interactions as an example. Comprehensive troubleshooting notes are provided to facilitate the adaptation of these protocols to different ligand-receptor systems. The methodology outlined for studying protein-ligand interactions will help to avoid common errors and misinterpretations of experimental results.
Der interdisziplinäre Sammelband eröffnet neue Perspektiven auf den Stil als bislang unterkonturierte literaturwissenschaftliche Leitkategorie unter transnationalen, wissens-, gattungs- und sprachgeschichtlichen Gesichtspunkten. Im 18. Jahrhundert zeichnet sich im Nachdenken über Schreibarten eine Neujustierung der Stilkategorie ab, die den Stil zur Reflexionsgröße für ästhetische Diskurse macht. Der Band sondiert die Pluralisierung, Historisierung und Individualisierung der Stilkategorie, die ihr neue literatur- und kulturtheoretische Anwendungsbereiche eröffnet. Die Bewegungen zwischen den Sprachen, Literaturen, Medien und semantischen Feldern erschließt die Publikation, indem sie europäische Vergleichshorizonte eröffnet und literatur- ebenso wie sprachwissenschaftliche Ansätze präsentiert. Damit leistet sie einen Beitrag zum Feld der komparatistisch ausgerichteten Germanistik, insbesondere der Literatur- und Kulturgeschichte des 18. Jahrhunderts und des europäischen Kulturtransfers.
The total charm-quark production cross section per unit of rapidity dσ(cc)/dy, and the fragmentation fractions of charm quarks to different charm-hadron species f(c → hc), are measured for the first time in p–Pb collisions at √sNN = 5.02 TeV at midrapidity (−0.96 < y < 0.04 in the centre-ofmass frame) using data collected by ALICE at the CERN LHC. The results are obtained based on all the available measurements of prompt production of ground-state charm-hadron species: D0, D+,D+s, and J/ψ mesons, and Λ+cand Ξ0cbaryons. The resulting cross section is dσ(cc)/dy = 219.6±6.3 (stat.)+10.5−11.8(syst.)+7.6−2.9(extr.)±5.4 (BR)±4.6 (lumi.)±19.5 (rapidity shape) +15.0 (Ω0c) mb, which is consistent with a binary scaling of pQCD calculations from pp ollisions. The measured fragmentation fractions are compatible with those measured in pp collisions at √s = 5.02 and 13 TeV, showing an increase in the relative production rates of charm baryons with respect to charm mesons in pp and p–Pb collisions compared with e+e − and e−p collisions. The pT-integrated nuclear modification factor of charm quarks, RpPb(cc) = 0.91±0.04 (stat.) +0.08 −0.09 (syst.) +0.04 −0.03 (extr.)±0.03 (lumi.), is found to be consistent with unity and with theoretical predictions including nuclear modifications of the parton distribution functions.
Natural Language Processing (NLP) for big data requires an efficient and sophisticated infrastructure to complete tasks both fast and correctly. Providing an intuitive and lightweight interaction with a framework that abstracts and simplifies complex tasks assists in reaching this goal. This bachelor thesis extends the NLP framework Docker Unified UIMA Interface (DUUI) by an API and a web-based graphical user interface to control and manage pipelines for automated analysis of large quantities of natural language. The extension aims to reduce the entry barrier into the field as well as to accelerate the creation and management of pipelines according to UIMA standards. Pipelines can be executed in the browser or using the web API directly and then monitored on a document level. The evaluation in usability and user experience indicates that the implementation benefits the framework by making its usage more user friendly, lightweight, and intuitive while also making the management of pipelines more efficient.
The e+e−→D+sDs1(2536)− and e+e−→D+sD∗s2(2573)− processes are studied using data samples collected with the BESIII detector at center-of-mass energies from 4.530 to 4.946~GeV. The absolute branching fractions of Ds1(2536)−→D¯∗0K− and D∗s2(2573)−→D¯0K− are measured for the first time to be (35.9±4.8±3.5)% and (37.4±3.1±4.6)%, respectively. The measurements are in tension with predictions based on the assumption that the Ds1(2536) and D∗s2(2573) are dominated by a bare cs¯ component. The e+e−→D+sDs1(2536)− and e+e−→D+sD∗s2(2573)− cross sections are measured, and a resonant structure at around 4.6~GeV with a width of 50~MeV is observed for the first time with a statistical significance of 15σ in the e+e−→D+sD∗s2(2573)− process. It could be the Y(4626) found by the Belle collaboration in the D+sDs1(2536)− final state, since they have similar masses and widths. There is also evidence for a structure at around 4.75~GeV in both processes.
Dass das ZfL ihn seit Kurzem als Senior Fellow führt, passt so gar nicht zu der ungestümen Neugierde und den kreativen Energien, die sich Detlev Schöttker nicht nur bewahrt hat, sondern die jüngst über der Beschäftigung mit einem neuen Gegenstand eine neue Qualität gewonnen haben. Instantan, vehement und bedingungslos hat er sich nach dem Umzug des ZfL nach Wilmersdorf in ein neues Forschungsprojekt weniger vertieft als gestürzt. Doch recht besehen ist es kein neuer Gegenstand, sondern es sind seine alten Bekannten, die ihm rund um den Fasanenplatz wiederbegegnen. Ein größeres Geschenk als diese Nachbarschaft hätte man ihm vielleicht nicht machen können: Die literarische und kulturelle Moderne entstand hier! Der Fasanenplatz ist ein 'Freilichtmuseum' der Moderne mit Hauptmann, Brecht, Benjamin und vielen anderen. Und Detlev Schöttker wäre nicht Detlev Schöttker, wenn er die Öffentlichkeit nicht sogleich über einige seiner Funde und Entdeckungen informiert hätte. In der FAZ sind bereits mehrere Artikel von ihm über die erstaunliche Bedeutung unseres Kiezes für die Moderne erschienen.
Das Fach "Allgemeine und vergleichende Literaturwissenschaft (AVL)" beschäftigt sich mit dem Vergleich von Literatur aus verschiedenen Kulturen, Epochen und Gattungen. Der Begriff "Komparatistik" wird weitgehend synonym verwendet, legt aber einen noch stärkeren Schwerpunkt auf den Vergleich als allgemeine epistemologische Praxis, vor dem der Fokus auf Literatur in den Hintergrund geraten kann. Die Geschichte des Fachs in Deutschland reicht bis in das 19. Jahrhundert zurück, in dem es - in etwa zeitgleich mit den "Nationalphilologien" (hier also Germanistik) - vor allem im Sinne einer vergleichenden Literaturgeschichte entstand. Während in den Anfängen Einflussforschung und Gattungsgeschichte im Vordergrund standen, hat das Fach im 20. Jahrhundert mehrere Ausweitungen erfahren. So zählen beispielsweise auch die allgemeine Literatur- und Kulturtheorie, die Ästhetik und der Vergleich von Literatur mit anderen Medien zu den Forschungsgebieten der AVL. Schon früh wurde der grenzüberschreitende Anspruch der AVL über den von J.W. Goethe lancierten Begriff "Weltliteratur" bestimmt. Entsprechend gehört auch die Frage, wie der Begriff historisch verstanden wurde und wie er im Zeitalter beschleunigter Globalisierung zu denken ist, zu den prominentesten Debatten im Rahmen der AVL. In der ursprünglichen Konzeption von Goethe war "Weltliteratur" als Dialog zwischen den Nationalliteraturen gedacht, wobei sowohl Rezeption (des kulturell Fremden) als auch Widerspiegelung (des Eigenen in der Rezeption durch Andere) im Vordergrund standen. Heute wird die eurozentrische Orientierung des Begriffs problematisiert, aber die Frage bleibt virulent, ob und wie Literatur aus verschiedenen Kulturen und Sprachen auf eine universelle Ebene gehoben werden kann, ohne ihre spezifischen Kontexte zu verlieren.
Als Carlo Ginzburg die These formulierte, dass die Geisteswissenschaften wie die Kriminalliteratur im sog. "Indizienparadigma" gründeten, hatte er mit Sherlock Holmes einen Detektiv vor Augen, der persönlich den Tatort besichtigte. Dort erhob er Spuren, kombinierte sie und kam in oftmals ingeniösen, aber auch höchst spekulativen Schlussfolgerungen zur Lösung seines Falls. Vor dem Hintergrund aktueller Entwicklungen in Forschung und Fahndung muss dieses materiell und empirisch grundierte "Indizienparadigma" jedoch einer Revision unterzogen werden. Denn seit der Privatdetektiv von "Kommissar Computer" Konkurrenz bekommen hat, haben sich die Investigationspraktiken grundlegend gewandelt: So können computergestützte Fahndungs- und Aufklärungsmethoden eine Besichtigung des Tatorts ersetzen, während algorithmische Wahrscheinlichkeitsrechnung vergangene wie zukünftige Fälle erhellt. Der vorliegende Sammelband mit Beiträgen aus der Literatur-, Medien- und Designwissenschaft untersucht, inwiefern solche "virtuellen Investigationen" in Literatur und Kunst der Gegenwart eine Revision des Indizienparadigmas einschließen - und inwiefern Begriffe der Virtualität bereits die Investigativarbeit im 19. Jahrhundert prägten.
The family of cubic noncentrosymmetric 3-4-3 compounds has become a fertile ground for the discovery of novel correlated metallic and insulating phases. Here, we report the synthesis of a new heavy fermion compound, Ce3Bi4Ni3. It is an isoelectronic analog of the prototypical Kondo insulator Ce3Bi4Pt3 and of the recently discovered Weyl-Kondo semimetal Ce3Bi4Pd3. In contrast to the volume-preserving Pt-Pd substitution, structural and chemical analyses reveal a positive chemical pressure effect in Ce3Bi4Ni3 relative to its heavier counterparts. Based on the results of electrical resistivity, Hall effect, magnetic susceptibility, and specific heat measurements, we identify an energy gap of 65-70 meV, about eight times larger than that in Ce3Bi4Pt3 and about 45 times larger than that of the Kondo-insulating background hosting the Weyl nodes in Ce3Bi4Pd3. We show that this gap as well as other physical properties do not evolve monotonically with increasing atomic number, i.e., in the sequence Ce3Bi4Ni3-Ce3Bi4Pd3-Ce3Bi4Pt3, but instead with increasing partial electronic density of states of the d orbitals at the Fermi energy. To understand under which condition topological states form in these materials is a topic for future studies.
Heterostructures of graphene in proximity to magnetic insulators open the possibility to investigate exotic states emerging from the interplay of magnetism, strain and charge transfer between the layers. Recent reports on the growth of self-integrated atomic wires of β-RuCl3 on graphite suggest these materials as versatile candidates to investigate these effects. Here we present detailed first principles calculations on the charge transfer and electronic structure of β-RuCl3/heterostructures and provide a comparison with the work function analysis of the related honeycomb family members α-RuX3 (X = Cl,Br,I). We find that proximity of the two layers leads to a hole-doped graphene and electron-doped RuX3 in all cases, which is sensitively dependent on the distance between the two layers. Furthermore, strain effects due to lattice mismatch control the magnetization which itself has a strong effect on the charge transfer. Charge accumulation in β-RuCl3 strongly drops away from the chain making such heterostructures suitable candidates for sharp interfacial junctions in graphene-based devices.
canning tunneling microscopy (STM) is perhaps the most promising way to detect the superconducting gap size and structure in the canonical unconventional superconductor Sr2RuO4 directly. However, in many cases, researchers have reported being unable to detect the gap at all in simple STM conductance measurements. Recently, an investigation of this issue on various local topographic structures on a Sr-terminated surface found that superconducting spectra appeared only in the region of small nanoscale canyons, corresponding to the removal of one RuO surface layer. Here, we analyze the electronic structure of various possible surface structures using first principles methods, and argue that bulk conditions favorable for superconductivity can be achieved when removal of the RuO layer suppresses the RuO4 octahedral rotation locally. We further propose alternative terminations to the most frequently reported Sr termination where superconductivity surfaces should be observed.
Motivated by recently reported magnetic-field induced topological phases in ultracold atoms and correlated Moiré materials, we investigate topological phase transitions in a minimal model consisting of interacting spinless fermions described by the Hofstadter model on a square lattice. For interacting lattice Hamiltonians in the presence of a commensurate magnetic flux it has been demonstrated that the quantized Hall conductivity is constrained by a Lieb-Schultz-Mattis (LSM)-type theorem due to magnetic translation symmetry. In this work, we revisit the validity of the theorem for such models and establish that a topological phase transition from a topological to a trivial insulating phase can be realized but must be accompanied by spontaneous magnetic translation symmetry breaking caused by charge ordering of the spinless fermions. To support our findings, the topological phase diagram for varying interaction strength is mapped out numerically with exact diagonalization for different flux quantum ratios and band fillings using symmetry indicators. We discuss our results in the context of the LSM-type theorem.
On the potential for GWAS with phenotypic population means and allele-frequency data (popGWAS)

This study explores the potential of a novel genome-wide association study (GWAS) approach for identifying loci underlying quantitative polygenic traits in natural populations. Extensive population genetic forward simulations demonstrate that the approach is generally effective for oligogenic and moderately polygenic traits and relatively insensitive to low heritability, but applicability is limited for highly polygenic architectures and pronounced population structure. The required sample size is moderate with very good results being obtained already for a few dozen populations scored. The method performs well in predicting population means even with a moderate false positive rate. When combined with machine learning for feature selection, this rate can be further reduced. The data efficiency of the method, particularly when using pooled sequencing, makes GWAS studies more accessible for research in biodiversity genomics. Overall, this study highlights the promise of this popGWAS approach for dissecting the genetic basis of complex traits in natural populations.
Motivated by the on-going discussion on the nature of magnetism in the quantum Ising chain CoNb2O6, we present a first-principles-based analysis of its exchange interactions by applying an \textit{ab initio} approach with additional modelling that accounts for various drawbacks of a purely density functional theory ansatz. With this method we are able to extract and understand the origin of the magnetic couplings under inclusion of all symmetry-allowed terms, and to resolve the conflicting model descriptions in CoNb2O6. We find that the twisted Kitaev chain and the transverse-field ferromagnetic Ising chain views are mutually compatible, although additional off-diagonal exchanges are necessary to provide a complete picture. We show that the dominant exchange interaction is a ligand-centered exchange process - involving the eg electrons -, which is rendered anisotropic by the low-symmetry crystal fields environments in CoNb2O6, giving rise to the dominant Ising exchange, while the smaller bond-dependent anisotropies are found to originate from d−d kinetic exchange processes involving the t2g electrons. We demonstrate the validity of our approach by comparing the predictions of the obtained low-energy model to measured THz and inelastic neutron scattering spectra.
he family of cubic noncentrosymmetric 3-4-3 compounds has become a fertile ground for the discovery of novel correlated metallic and insulating phases. Here, we report the synthesis of a new heavy fermion compound, Ce3Bi4Ni3. It is an isoelectronic analog of the prototypical Kondo insulator Ce3Bi4Pt3 and of the recently discovered Weyl-Kondo semimetal Ce3Bi4Pd3. In contrast to the volume-preserving Pt-Pd substitution, structural and chemical analyses reveal a positive chemical pressure effect in Ce3Bi4Ni3 relative to its heavier counterparts. Based on the results of electrical resistivity, Hall effect, magnetic susceptibility, and specific heat measurements, we identify an energy gap of 65–70 meV, about eight times larger than that in Ce3Bi4Pt3 and about 45 times larger than that of the Kondo-insulating background hosting the Weyl nodes in Ce3Bi4Pd3. We show that this gap as well as other physical properties do not evolve monotonically with increasing atomic number, i.e., in the sequence Ce3Bi4Ni3−Ce3Bi4Pd3−Ce3Bi4Pt3, but instead with increasing partial electronic density of states of the 𝑑 orbitals at the Fermi energy. This work opens the possibility to investigate the conditions under which topological states develop in this series of strongly correlated 3-4-3 materials.
Abstract
Seed harvesting from wild plant populations is key for ecological restoration, but may threaten the persistence of source populations. Consequently, several countries have set guidelines limiting the proportions of harvestable seeds. However, these guidelines are so far inconsistent, and they lack a solid empirical basis. Here, we use high-resolution data from 298 plant species to model the demographic consequences of seed harvesting. We find that the current guidelines do not protect populations of annuals and short-lived perennials, while they are overly restrictive for long-lived plants. We show that the maximum possible fraction of seed production – what can be harvested without compromising the long-term persistence of populations – is strongly related to the generation time of the target species. When harvesting every year, this safe seed fraction ranges from 80% in long-lived species to 2% in most annuals. Less frequent seed harvesting substantially increases the safe seed fraction: In the most vulnerable annual species, it is safe to harvest 5%, 10% or 30% of population seed production when harvesting every two, five or ten years, respectively. Our results provide a quantitative basis for seed harvesting legislations worldwide, based on species’ generation time and harvesting regime.
Significance The UN Decade on Ecosystem Restoration, 2021-2030, foresees upscaling restoration, and the demand for native seed is skyrocketing. Seeds for restoring native vegetation are often harvested in wild, but too intensive harvest can threaten the donor populations. Existing guidelines that set limits to wild seed harvest are mostly based on expert opinions, yet they commonly lack empirical basis and vary among regions in one order of magnitude. We show that the current guidelines urgently need to be reformulated, because they are overly restrictive in long-lived species, while they do not protect annual plants from extinction. Using matrix population models of nearly 300 plant species, we provide a quantitative basis for a new seed harvesting legislation world-wide.
Dynamic imaging of landmark organelles, such as nuclei, cell membrane, nuclear envelope, and lipid droplets enables image-based phenotyping of functional states of cells. Multispectral fluorescent imaging of landmark organelles requires labor-intensive labeling, limits throughput, and compromises cell health. Virtual staining of label-free images with deep neural networks is an emerging solution for this problem. Multiplexed imaging of cellular landmarks from scattered light and subsequent demultiplexing with virtual staining saves the light spectrum for imaging additional molecular reporters, photomanipulation, or other tasks. Published approaches for virtual staining of landmark organelles are fragile in the presence of nuisance variations in imaging, culture conditions, and cell types. This paper reports model training protocols for virtual staining of nuclei and membranes robust to label-free imaging parameters, cell states, and cell types. We developed a flexible and scalable convolutional architecture, named UNeXt2, for supervised training and self-supervised pre-training. The strategies we report here enable robust virtual staining of nuclei and cell membranes in multiple cell types, including neuromasts of zebrafish, across a range of imaging conditions. We assess the models by comparing the intensity, segmentations, and application-specific measurements obtained from virtually stained and experimentally stained nuclei and membranes. The models rescue the missing label, non-uniform expression of labels, and photobleaching. We share three pre-trained models, named VSCyto3D, VSCyto2D, and VSNeuromast, as well as VisCy, a PyTorch-based pipeline for training, inference, and deployment that leverages the modern OME-Zarr format.
Cyclin CLB2 mRNA localization and protein synthesis link cell cycle progression to bud growth

Clb2 is a conserved mitotic B-type cyclin, the levels of which are finely controlled to drive progression through the cell cycle. While it is known that CLB2 transcription and Clb2 protein degradation are important for precise control of its expression, it remains unclear whether the synthesis of Clb2 is also regulated. To address whether and how Clb2 expression levels respond to cell growth changes and adapt cell cycle progression, we combined single-cell and single-molecule imaging methods to measure CLB2 mRNA and protein expression throughout the Saccharomyces cerevisiae cell cycle. We found that the CLB2 mRNA was efficiently localized to the yeast bud as soon as this compartment was formed, but strikingly the Clb2 protein accumulated in the mother nucleus. The CLB2 mRNA localization in the yeast bud by the She2-3 complex did not control protein localization but rather promoted CLB2 translation. Moreover, CLB2 mRNA bud localization and protein synthesis were coupled and dependent on a single secondary structure -a ZIP code-located in the coding sequence. In a CLB2 ZIP code mutant, mRNA localization was impaired and Clb2 protein synthesis decreased, resulting in changes in cell cycle distribution and increased size of daughter cells at birth. Finally, while in WT cells the Clb2 protein concentration followed bud growth, this relationship was impaired in the ZIP code mutant. We propose that S. cerevisiae couples the control of CLB2 mRNA bud localization and protein synthesis to coordinate cell growth and cell cycle progression. This mechanism extends our knowledge of CLB2 expression regulation, and constitutes a novel function for mRNA localization.

Abstract
We study the impact of immigrant legalization on fiscal transfers from state to local governments in the United States, exploiting variation in legal status from the 1986 Immigration Reform and Control Act (IRCA). State governments allocate more resources to IRCA counties, an allocation that is responsive to the electoral incentives of the governor. Importantly, the effect emerges prior to the enfranchisement of the IRCA migrants and we argue it is driven by the IRCA’s capacity to politically empower already legal Hispanic migrants in mixed legal status communities. The IRCA increases turnout in large Hispanic communities as well as Hispanic political engagement, without detectably triggering anti-migrant sentiment.
Antimatter is a research topic of fundamental interest. Sufficient matter-antimatter asymmetry in the early Universe created the matter-dominated world today. The origin of this asymmetry is not completely understood to date. High-energy nuclear collisions create conditions similar to the Universe microseconds after the Big Bang, with comparable amounts of matter and antimatter. Much of the antimatter created escapes the rapidly expanding fireball without annihilation, making such collisions an effective experimental tool to create heavy antimatter nuclear objects and study their properties. In this paper, we report the first observation of the antimatter hypernucleus 4Λ¯H¯¯¯¯, composed of an Λ¯, an antiproton and two antineutrons. The discovery was made through its two-body decay after production in ultrarelativistic heavy-ion collisions by the STAR experiment at the Relativistic Heavy Ion Collider. In total, 15.6 candidate 4Λ¯H¯¯¯¯ antimatter hypernuclei are obtained with an estimated background count of 6.4. Lifetimes of the antihypernuclei 3Λ¯H¯¯¯¯ and 4Λ¯H¯¯¯¯ are measured and compared with the lifetimes of their corresponding hypernuclei, testing the symmetry between matter and antimatter. Various production yield ratios among (anti)hypernuclei and (anti)nuclei are also measured and compared with theoretical model predictions, shedding light on their production mechanism.
We report results on an elastic cross section measurement in proton-proton collisions at a center-of-mass energy s√=510 GeV, obtained with the Roman Pot setup of the STAR experiment at the Relativistic Heavy Ion Collider (RHIC). The elastic differential cross section is measured in the four-momentum transfer squared range 0.23≤−t≤0.67 GeV2. We find that a constant slope B does not fit the data in the aforementioned t range, and we obtain a much better fit using a second-order polynomial for B(t). The t dependence of B is determined using six subintervals of t in the STAR measured t range, and is in good agreement with the phenomenological models. The measured elastic differential cross section dσ/dt agrees well with the results obtained at s√=546 GeV for proton--antiproton collisions by the UA4 experiment. We also determine that the integrated elastic cross section within the STAR t-range is σfidel=462.1±0.9(stat.)±1.1(syst.)±11.6(scale) μb.

We measure triangular flow relative to the reaction plane at 3 GeV center-of-mass energy in Au+Au collisions at the BNL Relativistic Heavy Ion Collider. A significant v3 signal for protons is observed, which increases for higher rapidity, higher transverse momentum, and more peripheral collisions. The triangular flow is essentially rapidity-odd with a slope at mid-rapidity, dv3/dy|(y=0), opposite in sign compared to the slope for directed flow. No significant v3 signal is observed for charged pions and kaons. Comparisons with models suggest that a mean field potential is required to describe these results, and that the triangular shape of the participant nucleons is the result of stopping and nuclear geometry.
We report the first measurements of cumulants, up to 4th order, of deuteron number distributions and proton-deuteron correlations in Au+Au collisions recorded by the STAR experiment in phase-I of Beam Energy Scan (BES) program at the Relativistic Heavy Ion Collider. Deuteron cumulants, their ratios, and proton-deuteron mixed cumulants are presented for different collision centralities covering a range of center-of-mass energy per nucleon pair sNN−−−−√~=~7.7 to 200~GeV. It is found that the cumulant ratios at lower collision energies favor a canonical ensemble over a grand canonical ensemble in thermal models. An anti-correlation between proton and deuteron multiplicity is observed across all collision energies and centralities, consistent with the expectation from global baryon number conservation. The UrQMD model coupled with a phase-space coalescence mechanism qualitatively reproduces the collision-energy dependence of cumulant ratios and proton-deuteron correlations.
In heavy-ion collision experiments, the global collectivity of final-state particles can be quantified by anisotropic flow coefficients (vn). The first-order flow coefficient, also referred to as the directed flow (v1), describes the collective sideward motion of produced particles and nuclear fragments in heavy-ion collisions. It carries information on the very early stage of the collision, especially at large pseudorapidity (η), where it is believed to be generated during the nuclear passage time. Directed flow therefore probes the onset of bulk collective dynamics during thermalization, providing valuable experimental guidance to models of the pre-equilibrium stage. In 2018, the Event Plane Detector (EPD) was installed in STAR and used for the Beam Energy Scan phase-II (BES-II) data taking. The combination of EPD (2.1<|η|<5.1) and high-statistics BES-II data enables us to extend the v1 measurement to the forward and backward η regions. In this paper, we present the measurement of v1 over a wide η range in Au+Au collisions at √sNN= 19.6 and 27 GeV using the STAR EPD. The results of the analysis at √sNN=19.6 GeV exhibit excellent consistency with the previous PHOBOS measurement, while elevating the precision of the overall measurement. The increased precision of the measurement also revealed finer structures in heavy-ion collisions, including a potential observation of the first-order event-plane decorrelation. Multiple physics models were compared to the experimental results. Only a transport model and a three-fluid hybrid model can reproduce a sizable v1 at large η as was observed experimentally. The model comparison also indicates v1 at large η might be sensitive to the QGP phase transition.
In a Quark-Gluon Plasma (QGP), the fundamental building blocks of matter, quarks and gluons, are under extreme conditions of temperature and density. A QGP could exist in the early stages of the Universe, and in various objects and events in the cosmos. The thermodynamic and hydrodynamic properties of the QGP are described by Quantum Chromodynamics (QCD) and can be studied in heavy-ion collisions. Despite being a key thermodynamic parameter, the QGP temperature is still poorly known. Thermal lepton pairs (e+e− and μ+μ−) are ideal penetrating probes of the true temperature of the emitting source, since their invariant-mass spectra suffer neither from strong final-state interactions nor from blue-shift effects due to rapid expansion. Here we measure the QGP temperature using thermal e+e− production at the Relativistic Heavy Ion Collider (RHIC). The average temperature from the low-mass region (in-medium ρ0 vector-meson dominant) is (1.99±0.24)×1012 K, consistent with the chemical freeze-out temperature from statistical models and the phase transition temperature from LQCD. The average temperature from the intermediate mass region (above the ρ0 mass, QGP dominant) is significantly higher at (3.40±0.55)×1012 K. This work provides essential experimental thermodynamic measurements to map out the QCD phase diagram and understand the properties of matter under extreme conditions.
With the STAR experiment at RHIC, we characterize √sNN = 200 GeV p+Au collisions by event activity (EA) measured within the pseudorapidity range η∈[−5,−3.4] in the Au-going direction and report correlations between this EA and hard- and soft-scale particle production at mid-rapidity (η∈[−1,1]). At the soft scale, charged particle production in low-EA p+Au collisions is comparable to that in \pp collisions and increases monotonically with increasing EA. At the hard scale, we report measurements of high transverse momentum (pT) jets in events of different EAs. In contrast to the soft particle production, high-pT particle production and EA are found to be inversely related. To investigate whether this is a signal of jet quenching in high-EA events, we also report ratios of pT imbalance and azimuthal separation of dijets in high- and low-EA events. Within our measurement precision, no significant differences are observed, disfavoring the presence of jet quenching in the highest 30% EA p+Au collisions at √sNN = 200 GeV.
Atomic nuclei are self-organized, many-body quantum systems bound by strong nuclear forces within femtometer-scale space. These complex systems manifest a diverse set of shapes~, traditionally explored via non-invasive spectroscopic techniques at low energies. Their instantaneous shapes, obscured by long-timescale quantum fluctuations, are considered not directly observable at low energy. We introduce a complementary method, collective flow assisted nuclear shape imaging, to image the nuclear global shape by colliding them at ultrarelativistic speeds and analyzing the collective response of outgoing debris. This technique captures a collision-specific snapshot of the spatial matter distribution in the nuclei, which, through the hydrodynamic expansion, leaves imprints on the particle momentum distribution patterns observed in detectors. We benchmark this method in collisions of ground state Uranium-238 nuclei, known for its elongated, axial-symmetric shape. Our findings, while confirming an overall deformation broadly consistent with prior low-energy experiments, also indicate a small deviation from axial symmetry in the nuclear ground state. This approach marks a new way of imaging nuclei, especially those with uncertain shape characteristics, and refines initial conditions in high-energy nuclear collisions. It tackles the important issue of nuclear structure evolution across various energy scales.
We report a new measurement of transverse single-spin asymmetries for dijet production in collisions of polarized protons at s√ = 200 GeV. Correlations between the proton spin and the transverse momenta of its partons, each perpendicular to the proton momentum direction, are probed at high Q2 ≈160 GeV2. The associated Sivers observable ⟨kT⟩, the average parton transverse momentum, is extracted using simple kinematics. Nonzero Sivers effects are observed for the first time in dijets from proton-proton collisions, but only when the jets are sorted by their net charge, which enhances the u- or d-quark contributions to separate data samples. This also enables a simple kinematic approach for determination of the individual partonic contributions to the observed asymmetries.

Using a sample of (10087±44)×106 𝐽/𝜓 events, which is about 45 times larger than that was previously analyzed, a further investigation on the 𝐽/𝜓→𝛾3(𝜋+𝜋−) decay is performed. A significant distortion at 1.84 GeV/𝑐2 in the line shape of the 3(𝜋+𝜋−) invariant mass spectrum is observed for the first time, which could be resolved by two overlapping resonant structures, 𝑋(1840) and 𝑋(1880). The new state 𝑋(1880) is observed with a statistical significance larger than 10𝜎. The mass and width of 𝑋(1880) are determined to be 1882.1±1.7±0.7 MeV/𝑐2 and 30.7±5.5±2.4 MeV, respectively, which indicates the existence of a 𝑝¯ 𝑝 bound state.
Using data samples collected with the BESIII detector operating at the BEPCII storage ring, the cross section of the inclusive process e+e−→η+X, normalized by the total cross section of e+e−→hadrons, is measured at eight center-of-mass energy points from 2.0000 GeV to 3.6710 GeV. These are the first measurements with momentum dependence in this energy region. Our measurement shows a significant discrepancy from calculations with the existing fragmentation functions. To address this discrepancy, a new QCD analysis is performed at the next-to-next-to-leading order with hadron mass corrections and higher twist effects, which can explain both the established high-energy data and our measurements reasonably well.

Zum Jahreswechsel 2023/2024 gelang einer russischen Fernsehserie, was während Russlands Krieg gegen die Ukraine eigentlich unvorstellbar scheint: Innerhalb weniger Tage entwickelte sich "Ehrenwort eines Kerls. Blut auf dem Asphalt" ("Slowo pazana. Krow na asfalte", 2023) beiderseits der Schützengräben zur populärsten Serie des Jahres. Die Zuschauer- und Klickzahlen erreichten Rekordhöhen und der Titelsong "Pyjala" (dt. 'Glas') der tatarischen Band Aigel schaffte es an die Spitze diverser Hitparaden in beiden Ländern. In der Russischen Föderation war die Serie zwar mit der Altersgrenze "18+" versehen und nur bei den privaten Streamingdiensten Wink und START zu sehen. Doch schon während der Ausstrahlung der acht Folgen der ersten Staffel vom 9. November bis 21. Dezember 2023 verbreitete die Serie sich blitzschnell über Telegram und andere digitale Kanäle. Sätze wie "Kerle entschuldigen sich nicht" oder "Denk dran, du bist jetzt ein Kerl, du bist jetzt auf der Straße, und ringsherum sind Feinde" wurden zu geflügelten Worten. Pädagogen und Politikerinnen schlugen Alarm, als in der Presse Berichte auftauchten, die von durch die Serie inspirierten Schlägereien berichteten, und zwar sowohl in Russland als auch in der Ukraine. Bevor die letzten Folgen überhaupt ausgestrahlt worden waren, gab es auf "Ehrenwort eines Kerls" bereits in beiden Ländern ein breites Medienecho, wobei die Kritiken kontrovers ausfielen und von enthusiastischer Begeisterung bis zu hellem Entsetzen und kategorischer Ablehnung reichten. In der Ukraine kreiste die Diskussion vor allem um die Frage, ob die Fernsehserie allein schon deshalb gefährliche Kriegspropaganda sei, weil sie aus dem Feindesland kommt. In Russland erregte die vermeintliche Romantisierung der Verbrecherwelt Anstoß. Manche Kritiker deuteten die Serie aber auch als subversiven Zerrspiegel der militärischen Aggression. Was war das aber für ein populärkulturelles Werk, das für so viel Aufmerksamkeit und Aufregung sorgte?
Autophagy is an important degradation pathway mediating the engulfment of cellular material (cargo) into autophagosomes followed by degradation in autophagosomes.
Different stress stimuli, e.g. nutrient deprivation, oxidative stress or organelle damage, engage autophagy to maintain cellular homeostasis, recycle nutrients or remove damaged cell organelles. Autophagy not only degrades bulk cytoplasmic material but also selective autophagic cargo, for example lysosomes (lysophagy), mitochondria (mitophagy), ER (ER-phagy), lipid droplets (lipophagy), protein aggregates (aggrephagy) or pathogens (xenophagy). Selective autophagy pathways are regulated by selective autophagy receptors which bind to ubiquitinated cargo proteins and link them to LC3 on the autophagosomal membrane.
Ubiquitination is an essential post-translational modification controlling different cellular processes such as proteasomal and lysosomal degradation or innate immune signaling.
M1-linked (linear) poly-Ubiquitin (poly-Ub) chains are exclusively assembled by the E3 ligase linear ubiquitin chain assembly complex (LUBAC) and removed by the M1 poly-Ub-specific OTU domain-containing deubiquitinase with linear linkage specificity (OTULIN). In addition to key functions in innate immune signaling and nuclear factor-κB (NF-κB) activation, M1 ubiquitination is also implicated in the regulation of autophagy.
LUBAC and OTULIN control autophagy initiation and maturation and the autophagic clearance of invading bacteria via xenophagy. However, additional functions of LUBAC- and OTULIN-regulated M1 ubiquitination in autophagy are largely unknown and it also remains unexplored if LUBAC and OTULIN control other selective autophagy pathways in addition to xenophagy. This study aimed to unravel the role of LUBAC- and OTULIN-controlled M1 ubiquitination in bulk and selective autophagy in more detail.
In this study, characterization of OTULIN-depleted MZ-54 glioblastoma (GBM) cells revealed that OTULIN deficiency results in enhanced LC3 lipidation in response to autophagy induction and upon blockade of late stage autophagy with Bafilomycin A1 (BafA1). Furthermore, electron microscopy analysis showed that OTULIN-deficient cells have an increased number of degradative compartments (DGCs), confirming enhanced autophagy activity upon loss of OTULIN. APEX2-based autophagosome content profiling identified various OTULIN-dependent autophagy cargo proteins. Among these were the autophagy receptor TAX1BP1 which regulates different forms of selective autophagy (e.g. lysophagy, aggrephagy) and the glycan-binding protein galectin-3 which serves key functions in lysophagy, suggesting a role of OTULIN and M1 poly-Ub in the regulation of aggrephagy and lysophagy.

To study aggrephagy, protein aggregation was induced with puromycin which causes premature termination of translation and accumulation of defective ribosomal products (DRiPs). Loss of OTULIN increased the number of M1 poly-Ub-positive foci and insoluble proteins and reduced the levels of soluble TAX1BP1 and p62 in response to puromycin-induced proteotoxic stress.
Intriguingly, upon induction of lysosomal membrane permeabilization (LMP) with the lysosomotropic drug L-Leucyl-L-Leucine methyl ester (LLOMe), M1 poly-Ub strongly accumulated at damaged lysosomes and colocalized with TAX1BP1- and galectin-3-positive puncta. M1 poly-Ub-modified lysosomes formed a platform for NF-κB essential modulator (NEMO) and inhibitor of κB (IκB) kinase (IKK) complex recruitment and local NF-κB activation in a K63 poly-Ub- and OTULIN-dependent manner. Furthermore, inhibition of lysosomal degradation enhanced LLOMe-induced cell death, suggesting pro-survival functions of lysophagy following LMP. Enrichment of M1 poly-Ub at damaged lysosomes was also observed in human dopaminergic neurons and in primary mouse embryonic cortical neurons, confirming the importance of M1 poly-Ub in the response to lysosomal damage.
Together, these results identify OTULIN as a negative regulator of autophagy induction and the autophagic flux and reveal OTULIN-dependent autophagy cargo proteins.
Furthermore, this study uncovers novel and important roles of M1 poly-Ub in the response to lysosomal damage and local NF-κB activation at damaged lysosomes.
Das partizipative Forschungsprojekt "Allein aber vernetzt? Digitale (Un)gleichheiten und soziale Netzwerke bei alleinlebenden Menschen" beschäftigte sich von 2021-2024 mit Alleinlebenden im Alter, ihren Beziehungsnetzwerken und der Frage welche Rolle digitale Geräte und die Digitalisierung aller Lebensbereiche in diesem Zusammenhang spielen. Durch die Zusammenarbeit mit einer Gruppe von Ko-Forschenden in Frankfurt und Egelsbach stand die Perspektive der Gruppe im Mittelpunkt.
Im vorliegenden White Paper wird das Projekt sowie zentrale Ergebnisse und die erarbeiteten Handlungsempfehlungen der deutschen Teilstudie vorgestellt. Diese wurde vom BMBF gefördert und ist Teil des europäischen Verbundprojektes "EQualCare - Alone but connected? Digital (in)equalities in care work and generational relationships among older people living alone" der Joint Programming Initiative (JPI) “More Years Better Lives”.
An independent Taiwanese lineage of powdery mildew on the endemic host species Koelreuteria henryi

Background: Powdery mildews (Erysiphaceae, Ascomycota) are common plant disease agents and also cause stress for forest and fruit trees worldwide as well as in Taiwan. The powdery mildew Erysiphe bulbouncinula on Koelreuteria host trees was considered an endemic species in China. While in China the host was K. paniculata and only the teleomorph stage found, the anamorph and the teleomorph were both recorded for the host in Taiwan, K. henryi. We aimed to clarify the relationship of the powdery mildews recorded under E. bulbouncinula with an apparently disjunct distribution.
Results: Specimens of powdery mildew on K. henryi from Taiwan were characterized based on the anamorph morphology and DNA sequences. They revealed a new record of Sawadaea koelreuteriae for this host species and Taiwan and a new species of Erysiphe, E. formosana, sister to E. bulbouncinula from China.
Conclusions: In Erysiphe on Koelreuteria hosts, speciation of plant parasitic fungi seems to be correlated with disjunct host and geographic distribution possibly shaped by extinction of potential host species which are known only as fossils. Two of the three extant East Asian species of Koelreuteria are now known as hosts of specific Erysiphe species. We may predict a further not yet discovered Erysiphe species on the third East Asian species, K. bipinnata, in South and Southwest China. In the speciation in Sawadaea, the extinction events in Koelreuteria can be excluded from being involved.
Das Schilddrüsenkarzinom (SK) ist die häufigste bösartige endokrine Tumorerkrankung. Während das nicht-metastasierte und nicht-mutierte papilläre Schilddrüsenkarzinom (PSK) und das follikuläre Schilddrüsenkarzinom (FSK) eine gute Heilungschance aufweisen, zeigen die mutierten und metastasierten Varianten des PSK und FSK sowie das anaplastische Schilddrüsenkarzinom (ASK) weiterhin eine schlechte Prognose. Die Entwicklung von Therapieresistenzen stellen hierbei ein Hauptproblem in der Behandlung des fortgeschrittenen Schilddrüsenkarzinoms dar.
In den letzten Jahren wurden in Studien zunehmend Tumor-initiierende Zellen (TIZ) beschrieben, welche eine kleine Subpopulation von Zellen mit der Fähigkeit zur Selbsterneuerung, Tumorinitiierung und Entwicklung von Therapieresistenzen von Tumoren darstellen. Die Existenz von TIZ wurde auch im SK nachgewiesen. Ein entscheidender Faktor für die Persistenz von TIZ ist die Hypoxie, welche über eine Veränderung des Tumormikromilieus und des Zellmetabolismus zur Entstehung von Therapieresistenzen beiträgt. Ein durch Hypoxie hochreguliertes Enzym ist die Carboanhydrase IX (CAIX). CAIX wird hauptsächlich von Tumorzellen exprimiert und katalysiert die Reaktion von Kohlendioxid zu Bicarbonat und einem Proton und trägt damit zur Säurepufferung der Tumorzelle bei. CAIX stellt somit einen entscheidenden Faktor für das Überleben von Tumorzellen in einem hypoxischen Milieu dar. Des Weiteren ist eine erhöhte Expression von CAIX mit einem schlechten Patienten-Outcome assoziiert, wie z.B. im Brustkrebs. Diese Eigenschaften machen CAIX zu einem attraktiven Angriffspunkt einer zielgerichteten Tumortherapie. Die vorliegende Studie hat zum Ziel, die Expression von CAIX sowie dessen biologische Rolle im Schilddrüsenkarzinom näher zu untersuchen.
Hierzu wurden Proben von 114 SK-Patienten immunhistochemisch auf eine CAIX-Expression untersucht und mit tumorfreiem Schilddrüsengewebe verglichen. Hierbei waren unterschiedliche SK-Subtypen vertreten. Für eine weitere Validierung der Expressionsdaten erfolgte die Auswertung eines Datasets von „The Cancer Genome Atlas“ (TCGA) mithilfe von cBioportal. Da die Hypoxie ein wichtiger Faktor für die Persistenz von TIZ ist, wurde die CAIX-Expression in Tumorsphären, ein in vitro Nachweis von TIZ-Aktivität, mittels der Durchflusszytometrie bestimmt und mit der CAIX-Expression von Monolayern verglichen. Als SK-Zelllinien wurden BCPAP (PSK), FTC 133 (FSK) und 8505 C (ASK) verwendet. Anschließend wurde mithilfe der Polymerasekettenreaktion und Immunofluoreszenzfärbung untersucht, ob eine CAIX-Expression in den Tumorsphären mit der Expression von bereits bekannten Stammzellmarkern, u.a. NANOG, assoziiert ist. Die Unterschiede der CAIX-Expression, nach Inkubation der Monolayer jeweils in Normoxie und Hypoxie, wurden mittels Durchflusszytometrie bestimmt. Mithilfe eines genetischen CAIX-Knockdowns sowie einer pharmakologischen Inhibition mit dem CAIX-Inhibitor Methazolamid (MZM) wurde die Tumorzellproliferation und -Sphärenbildung unter Normoxie und Hypoxie bestimmt. Zusätzlich wurde der Einfluss von MZM auf die Apoptose und den Zellzyklus untersucht.
Immunhistochemische Färbungen der Gewebeproben von SK-Patienten zeigten, dass die CAIX-Expression sowohl im PSK und FSK als auch im ASK im Vergleich zum tumorfreien Schilddrüsengewebe erhöht war. Des Weiteren zeigte die klinisch-pathologische Datenanalyse, dass eine erhöhte CAIX-Expression mit dem Auftreten von Lymphknotenmetastasen im differenzierten SK assoziiert war. Auch die Analyse des TCGA-Datasets bestätigte, dass eine erhöhte Expression der CAIX-mRNA mit einem fortgeschrittenen Tumorstadium, Fernmetastasen und mit einem kürzeren Gesamt-Überleben von SK-Patienten korrelierte. Die weiteren funktionellen in vitro Untersuchungen ergaben, dass die CAIX-Expression in den Tumorsphären im Vergleich zu Monolayern erhöht und mit einer erhöhten Expression von Stammzellmarkern assoziiert war. Ein genetischer CAIX-Knockdown und eine CAIX-Inhibition mit MZM führten über eine Induktion der Apoptose und eines Zellzyklusarrests zu einer verminderten Tumorzellproliferation und Sphärenbildung.
Zusammenfassend deuten die Ergebnisse darauf hin, dass CAIX ein vielversprechendes Zielmolekül für eine gezielte Tumortherapie des fortgeschrittenen SK ist. Um diese Hypothese bestätigen zu können, sind jedoch weitere prospektive Analysen von Patientenproben sowie funktionelle in vivo Untersuchungen am SK nötig.
Muller's ratchet, in its prototype version, models a haploid, asexual population whose size~N is constant over the generations. Slightly deleterious mutations are acquired along the lineages at a constant rate, and individuals carrying less mutations have a selective advantage. The classical variant considers {\it fitness proportional} selection, but other fitness schemes are conceivable as well. Inspired by the work of Etheridge et al. ([EPW09]) we propose a parameter scaling which fits well to the ``near-critical'' regime that was in the focus of [EPW09] (and in which the mutation-selection ratio diverges logarithmically as N→∞). Using a Moran model, we investigate the``rule of thumb'' given in [EPW09] for the click rate of the ``classical ratchet'' by putting it into the context of new results on the long-time evolution of the size of the best class of the ratchet with (binary) tournament selection, which (other than that of the classical ratchet) follows an autonomous dynamics up to the time of its extinction. In [GSW23] it was discovered that the tournament ratchet has a hierarchy of dual processes which can be constructed on top of an Ancestral Selection graph with a Poisson decoration. For a regime in which the mutation/selection-ratio remains bounded away from 1, this was used in [GSW23] to reveal the asymptotics of the click rates as well as that of the type frequency profile between clicks. We will describe how these ideas can be extended to the near-critical regime in which the mutation-selection ratio of the tournament ratchet converges to 1 as N→∞.

Finanzielle Armut prägt Mobilitätspraktiken und kann dabei zum Prozess von mobilitätsbezogener sozialer Exklusion beitragen. Zu den Personen, deren Armutsrisiko besonders hoch ist, zählen in Deutschland Haushalte mit Kindern, insbesondere Alleinerziehende. Ältere Menschen haben nicht die höchste Armutsgefährdung, jedoch besteht bei ihnen das Risiko von Verharrung in Armut, da die Möglichkeiten, die finanzielle Situation aus eigener Kraft zu ändern mit zunehmendem Alter sinken.
Um ein tieferes Verständnis davon zu erhalten, wie finanzielle Armut die Mobilitätspraktiken und soziale Teilhabe von Haushalten mit Kindern sowie älteren Menschen prägt, wurden mit diesen beiden Personengruppen problemzentrierte Interviews in Ronnenberg (Region Hannover) geführt und analysiert. Die Ergebnisse belegen, dass, wenngleich alle Befragten mit ähnlich geringen finanziellen Ressourcen haushalten und Verzicht sowie Abwägungsprozesse notwendig sind, sich ihre Mobilitätspraktiken und Alltagsbewältigungsstrategien unterscheiden, was sich in zwei Typologien widerspiegelt. Erstens, eine Typologie der Mobilitätspraktiken von Haushalten mit Kindern: (i) autozentriert, (ii) autoreduziert, (iii) ÖPNV-orientiert und (iv) nichtmotorisiert. Zweitens, eine Typologie älterer Menschen anhand ihrer Mobilitätspraktiken: (i) aktive ältere Menschen mit vielseitigen sozialen Interaktionen, (ii) nachbarschaftsorientierte ältere Menschen mit lokalen Kontakten und (iii) ältere Menschen, die überwiegend zu Hause sind und wenig soziale Kontakte haben.
Um herauszufinden, inwiefern mobilitätsbezogene Barrieren der sozialen Teilhabe reduziert werden können, wurden fünf Maßnahmen bezüglich ihrer Wirkung auf die Mobilitätspraktiken einkommensarmer Haushalte mit Kindern untersucht: einerseits die Wirkung des 9-Euro-Tickets anhand von problemzentrierten Interviews mit einkommensarmen Haushalten mit Kindern, andererseits anhand von Expert:inneninterviews die Wirkung von Radlernkursen für Frauen mit Migrationshintergrund, eines Mietertickets, eines Quartierstickets und der Verbesserung der Nahraum- und Aufenthaltsqualität am Beispiel von Tempo 30. Die Ergebnisse zum 9-Euro-Ticket belegen, dass ein erschwingliches ÖPNV-Ticket erheblich zur Reduzierung mobilitätsbezogener Barrieren der sozialen Teilhabe im Armutskontext beiträgt. Die Expert:inneninterviews zeigen auf, dass eine Förderung des Umweltverbunds zielführend ist, um zu einer sozial-ökologischen Verkehrswende beizutragen und insbesondere Maßnahmenbündel Wirkung auf die Reduzierung von mobilitätsbezogenen Barrieren der sozialen Teilhabe entfalten.
Die Erkenntnisse dieser Dissertation ergänzen den wissenschaftlichen Forschungstand um ein tiefergehendes Verständnis der Wirkung von finanzieller Armut auf die Mobilitätspraktiken und soziale Teilhabe von Haushalten mit Kindern und älteren Menschen und helfen dabei, Maßnahmen zur Reduzierung mobilitätsbezogener Barrieren der sozialen Teilhabe zu konzipieren und umzusetzen.
Der Artikel bietet eine Analyse von John A. Williams' Roman "Clifford's Blues" (1999), der in der Form eines fiktiven, von einem afroamerikanischen Jazz-Musiker geschriebenen Tagebuchs von der Inhaftierung im Konzentrationslager Dachau erzählt. Der Roman lässt sich nicht nur als mit den Mitteln der Fiktion arbeitender Beitrag zur Geschichte der Verfolgung Schwarzer Menschen im Nationalsozialismus verstehen, sondern auch als selbst transatlantisches, vermeintlich historisches Zeitzeugnis, das aus einer ungewöhnlichen Perspektive die Thematik eines unsteten Archivs verflochtener Gewalt- und Exilerfahrungen beleuchtet. Das Augenmerk der Analyse liegt einerseits auf den Chancen eines solchen Archivs: Der Roman zeigt Möglichkeiten diasporischer Gemeinschaftsbildung und beschreibt die transatlantische Perspektive des Protagonisten als Ressource für das Erkennen von Kontinuitäten der Unterdrückung wie auch für das Ableiten von alltäglichen Widerstandsstrategien. Andererseits werden ausgehend vom Kunstgriff der Herausgeberfiktion und von Dokumenten aus dem Nachlass Risiken und Widersprüche des hier entworfenen Archivs diskutiert. Es wird gezeigt, dass dieses zwar auf Transnationalität zielt, jedoch teilweise der Rückbindung an ein lokales Moment bedarf.
Der Natrium-abhängige Kaliumkanal Slack (KNa1.1, Slo2.2, KCNT1) nimmt eine Schlüsselrolle in der Regulation neuronaler Erregbarkeit ein, indem er die Ausbildung und Feuerungsfrequenz von Aktionspotentialen kontrolliert. Sowohl in Mäusen als auch in Menschen wird Slack besonders hoch in nicht-peptidergen C-Faser-Neuronen exprimiert. Wissenschaftliche Erkenntnisse der letzten Jahre konnten die Beteiligung von Slack-Kanälen in der Signalverarbeitung neuropathischer Schmerzen, aber auch in verschiedenen Arten von Pruritus, feststellen. Dabei zeigen Slack-defiziente Mäuse ein verstärktes mechanisches Schmerzverhalten nach einer peripheren Nervenverletzung und ein erhöhtes Kratzverhalten in akuten Juckreiz-Modellen. Das als Slack-Aktivator identifizierte trizyklische Neuroleptikum Loxapin zeigt sowohl analgetische als auch antipruritische Effekte in Mäusen, jedoch ist sein klinischer Einsatz auf Grund schwerwiegender antipsychotischer Nebenwirkungen limitiert. Basierend auf Loxapins Leitstruktur wurden daher in dieser Arbeit neue Slack-Aktivatoren mit einem verbesserten pharmakologischen Profil designed und ihr Potential für die Therapie von Schmerzen sowie akutem und chronischem Pruritus in vivo untersucht.
The process 𝑒+𝑒−→Σ+¯Σ− is studied from threshold up to 3.04 GeV/𝑐2 via the initial-state radiation technique using data with an integrated luminosity of 12.0 fb−1, collected at center-of-mass energies between 3.773 and 4.258 GeV with the BESIII detector at the BEPCII collider. The pair production cross sections and the effective form factors of Σ are measured in eleven Σ+¯Σ− invariant mass intervals from threshold to 3.04 GeV/𝑐2. The results are consistent with the previous results from Belle and BESIII. Furthermore, the branching fractions of the decays 𝐽/𝜓→Σ+¯Σ− and 𝜓(3686)→Σ+¯Σ− are determined and the obtained results are consistent with the previous results of BESIII.
Bounding Dark Energy from the SPARC rotation curves: Data driven probe for galaxy virialization

Dark Energy (DE) acts as a repulsive force that opposes gravitational attraction. Assuming galaxies maintain a steady state over extended periods, the estimated upper bound on DE studies its resistance to the attractive gravitational force from dark matter. Using the SPARC dataset, we fit the Navarro-Frenk-White (NFW) and Hernquist models to identify the most suitable galaxies for these models. Introducing the presence of DE in these galaxies helps establish the upper limit on its repulsive force. This upper bound on DE sits around ρ(<Λ)∼10−25~kg/m3, only two orders of magnitude higher than the one measured by Planck. We discuss the conditions for detecting DE in different systems and show the consistency of the upper bound from galaxies to other systems. The upper bound is of the same order of magnitude as ρ200=200ρc for both dark matter profiles. We also address the implications for future measurements on that upper bound and the condition for detecting the impact of Λ on galactic scales.
The production cross section of inclusive isolated photons has been measured by the ALICE experiment at the CERN LHC in pp collisions at centre-of-momentum energy of s√=13 TeV collected during the LHC Run 2 data-taking period. The measurement is performed by combining the measurements of the electromagnetic calorimeter EMCal and the central tracking detectors ITS and TPC, covering a pseudorapidity range of |ηγ|<0.67 and a transverse momentum range of 7<pγT<200 GeV/c. The result extends to lower pγT and xγT=2pγT/s√ ranges, the lowest xγT of any isolated photon measurements to date, extending significantly those measured by the ATLAS and CMS experiments towards lower pγT at the same collision energy with a small overlap between the measurements. The measurement is compared with next-to-leading order perturbative QCD calculations and the results from the ATLAS and CMS experiments as well as with measurements at other collision energies. The measurement and theory prediction are in agreement with each other within the experimental and theoretical uncertainties.
Inorganic phosphate is one of the most abundant and essential nutrients in living organisms. It plays an indispensable role in energy metabolism and serves as a building block for major cellular components such as the backbones of DNA and RNA, headgroups of phospholipids and in posttranslational modifcations of many proteins. Disturbances in cellular phosphate homeostasis have a detrimental effect on the viability of cells. There- fore, both the import and export of phosphate is strictly regulated in eukaryotic cells. In the eukaryotic model organism Saccharomyces cerevisiae, the uptake of phosphate is carried out either by transporters with high affinity or by transporters with low affinity, depending on the cytosolic phosphate concentration. While structures are available for homologues of the high-affinity transporters, no structures of low-affinity transporters have been solved so far. Interestingly, only the low-affinity transporters have a regulatory SPX domain, which is found in various proteins involved in phosphate homeostasis.
In this work, structures of Pho90 from Saccharomyces cerevisiae, a low-affinity phosphate transporter, were solved by cryo-EM, providing insights into its transport mechanism. The dimeric structure resembles the structures of proteins of the divalent anion symporter superfamily (DASS) and of mammalian transporters of the solute carrier 13 (SLC13) family. The transmembrane domain of each protomer consists of 13 helical elements and can be subdivided into scaffold and transport domains. The structure of ScPho90 in the presence of phosphate shows the phosphate binding site within the transporter domain in an outward-open conformation with a bound phosphate ion and two sodium ions. In the absence of phosphate, an asymmetric dimer structure was determined, with one protomer adopting an inward-open conformation. While the dimer contact and the scaffold domain are identical in both conformations, the transport domain is rotated by about 30° and shifted by 11 Å towards the cytoplasmic side, leading to the accessibility of the binding pocket from the cytoplasm. Based on these findings and by comparison with known structures, a phosphate transport mechanism is proposed in the present work that involves substrate binding on the extracellular side, conformational change by a rigid-body motion of the transport domain, in an "elevator-like" motion, and substrate release into the cytoplasm. The regulatory SPX domain is not well resolved in the ScPho90 structures, so that no direct conclusions were drawn about its regulatory mechanism. The findings provide new insights into the function and mechanism of eukaryotic low-affinity phosphate transporters.
While eukaryotic cells express various phosphate import proteins, most eukaryotes have only a single highly conserved and essential phosphate exporter. These exporters show no sequence homology to other transporters of known structure, but also possess a regulatory SPX domain. In this work, the structural basis for eukaryotic phosphate export is investigated by elucidating the structures of the homologous phosphate exporters Syg1 from Saccharomyces cerevisiae and Xpr1 from Homo sapiens, using cryo-EM. The structures of ScSyg1 and HsXpr1 show a conserved homodimeric structure and the transmembrane part of each protomer consists of 10 TM helices. Helix TM1 establishes the dimer contact by means of a glycine zipper motif, which is a known oligomerization motif. Helices TM2-5 form a hydrophobic pocket that has density for a lipid molecule. Whether the lipid binding into the hydrophobic pocket has an allosteric effect on the phosphate export activity or only serves protein stabilization is not known. Helices TM5-10 form a six-helix bundle, which constitutes a putative phosphate translocation pathway in its center. This bundle is formed by the protein sequence annotated as EXS domain.
The respective phosphate translocation pathways of ScSyg1 and HsXpr1 show structural differences. While the translocation pathway in HsXpr1 is accessible from the cytoplasm, in ScSyg1 it is closed by a large loop of the SPX domain. Interestingly, this loop is not conserved in higher eukaryotes and is therefore not present in HsXpr1. Another difference are distinct conformations of helix TM9. In ScSyg1, TM9 adopts a kinked conformation, which results in the translocation pathway being open to the extracellular side. In contrast, TM9 adopts a straight conformation in HsXpr1, resulting in the placement of a highly conserved tryptophane residue in the middle of the translocation pathway. As a result, the translocation pathway in HsXpr1 is closed to the extracellular side.
Einleitung: Die Zentrale Notaufnahme (ZNA) stellt eine Schnittstelle zwischen prä- und innerklinischer Versorgung dar. Das Spektrum der Krankheitsbilder erstreckt sich über jegliche Fachrichtungen und variiert von harmlosen Banalitäten bis zu akuten Notfällen. Eine sichere und suffiziente Primärversorgung ist die Basis eines qualitativ-hochwertigen Gesundheitssystems.2 Verspätete oder falsche Diagnosen in der ZNA sind mit 10-30 % keine Seltenheit.
Dies ist nicht nur für den individuellen Patienten belastend, es bedeutet auch einen zusätzlichen Ressourcenverbrauch und eine finanzielle Belastung für das Gesundheitssystem. Clinical Decision Support Systems (CDSS) haben das Potenzial, sowohl professionelle Anwender als auch Laien bei ihrer Diagnosefindung zu unterstützen. Fragestellung: Ziel der Arbeit ist es herauszufinden, welchen Einfluss der Diagnosezeitpunkt auf das Outcome von Patienten mit Abdominalschmerzen in der ZNA hat und inwiefern ein CDSS das Potenzial hat, die genannten Punkte zu beeinflussen.
Methoden: Es handelte sich um eine prospektive, doppelt verblindete Beobachtungsstudie. Patienten, die sich mit Abdominalschmerzen in der Notaufnahme vorstellten, gaben ihre Symptome in die Ada-App ein. Sowohl die Diagnosevorschläge der App als auch die Verdachtsdiagnosen des behandelnden Arztes wurden dokumentiert und verglichen. Weitere erhobene Parameter waren die Verwendung von apparativer Diagnostik, die vergangene Zeit bis zur endgültigen Diagnosestellung, das Auftreten von Komplikationen, die Komorbidität und Mortalität sowie die Länge des Krankenhausaufenthalts. Das Follow-Up erfolgte zu verschiedenen Zeitpunkten bis zu Tag 90. Für die Analyse wurden die 450 Patienten anhand des Zeitpunkts ihrer Diagnosestellung in Gruppen "früh" (Tag 0) und "spät" (Tag 1-24) eingeteilt.
Ergebnisse: Im Vergleich zur „frühen“ Gruppe, hatte die Gruppe der „spät“ diagnostizierten Patienten einen höheren Anteil von Männern (45.2% (n=168/372) versus 60.3 % (n=47/78); p=0.018), im Schnitt einen höheren Charlson Comorbidity Index (0.7 versus 1.1; p=0.045) und im Schnitt einen höheren RAI-C Score (8.06 versus 9.9; p<0.001) am Tag ihrer Vorstellung. Bei den „spät“ diagnostizierten blieben weniger Patienten komplikationsfrei (57.0 % 49 (n=212/372) versus 17.9% (n=14/78); p<0.001), es traten mehr Major-Komplikationen auf (8.9% (n=33/372) versus 17.9% (n=14/78); p=0.024), analog dazu war der Comprehensive Complication Index höher (13.1 versus 25.6; p<0.001) und sie verweilten länger im Krankenhaus (2.6 Tage versus 6.7 Tage; p<0.001). Zudem nahmen sie signifikant mehr apparative Diagnostik in Anspruch.
Die behandelnden Ärzte konnten in 82.6% der Fälle (n=372/450) am Tag der Vorstellung die korrekte Diagnose stellen. Die Ada-App konnte in insgesamt 52% der Fälle (n=234/450) die korrekte Diagnose unter ihren Diagnoseverschlägen nennen.
Schlussfolgerung: Multimorbide Patienten scheinen anfälliger zu sein für falsche und verspätete Diagnosen. Ein später Diagnosezeitpunkt korreliert mit der vermehrten Nutzung apparativer Diagnostik, einem komplikationsreicheren Verlauf, einer höheren Komorbidität und einem längeren Krankenhausaufenthalt.
Die Ada-App ist den Ärzten zwar unterlegen, dennoch ist Potenzial vorhanden.
Für Ärzte kann die Ada-App eine Unterstützung im Rahmen der Diagnosefindung darstellen. Neben der Ressourcenentlastung kann sie vor Fehlannahmen und frühzeitigen Schlussfolgerungen schützen und auf weitere mögliche Differentialdiagnosen hinweisen. Die Ada-App stellt für Laien mit Sicherheit eine Weiterentwicklung gegenüber der simplen Symptomsuche über das Internet dar, dennoch sollten weitere Studien den Nutzen und die Sicherheit der App überprüfen.
In Deutschland leidet ca. jeder zehnte Mensch über 40 Jahren an einer chronischen Einschränkung seiner Nierenfunktion. Nicht wenige davon sind im Laufe der Erkrankung auf eine Nierenersatztherapie angewiesen. Die Ursachen für eine Nierenschädigung sind vielfältig. Als neuartiger und vielversprechender Therapieansatz werden aktuell Mesenchymale Stamm-/Stromazellen (MSC) als Therapeutikum für diverse Nierenerkrankungen getestet. Erste Ergebnisse klinischer Phase-I-Studien zeigen, dass MSC sicher als Immunsuppressivum nach Nierentransplantation angewendet werden können. Auch für weitere Erkrankungen der Niere sind erste klinische Studien am Laufen. MSC gelten als regenerativ, immunsupprimierend und antientzündlich. Dennoch gibt es noch einige Limitationen. Nach der Transplantation der Zellen ist das Wachstum der Zellen oft eingeschränkt und es kommt zur vermehrten Apoptose. Auch wird immer wieder ein paradoxes und entzündungsförderndes Verhalten der MSC am Wirkort beobachtet. Ein wichtiger Lösungsansatz ist eine in vitro Vorbehandlung der MSC zur Modulierung der zellulären Eigenschaften. In dieser Arbeit wurden drei Substanzen und Arzneimittel auf ihre Fähigkeit untersucht, die entzündungshemmenden Eigenschaften der MSC zu verbessern und die entzündungsfördernden zu unterdrücken. Der Fokus lag hierbei auf dem Arzneimittel Niclosamid und den beiden bisher noch nicht zugelassenen Substanzen Berberin und Gedunin, die alle in vitro und in vivo bereits erste vielversprechende antientzündliche Wirkungen bewiesen haben. Für diese Arbeit wurden MSC aus Fettgewebe isoliert (ASC) und mit LPS oder einem Cytokin-Mix (Mischung aus TNF-α, IFN-γ und IL-1β) sowie den drei Substanzen stimuliert. Untersucht wurden im Anschluss die mRNA-Expressionen der gängigsten proinflammatorischen (TNF-α, IL-6, TLR-4, ICAM-1, HLA-DR) und antiinflammatorischen Marker (IDO, IL-10), sowie mittels ELISA die Protein-Freisetzung von IL-6 und IL-8. Die vielversprechendsten Ergebnisse ließen sich durch Berberin hervorrufen. Hier zeigte sich eine deutliche Senkung der IL-8-Konzentration im ELISA. Die Anwendung des Gedunin hatte keine signifikante Wirkung auf die ASC. Niclosamid hingegen scheint widererwarten sogar entzündungsfördernd zu wirken, in dem es die IL-6-, ICAM-1-mRNA-Expression steigerte und die IDO-mRNA-Expression absenkte. Unter den drei getesteten Subtanzen hat Berberin die beste Wirkung bewiesen. Nach weiterer Testung könnte eine Anwendung mit Berberin als in vitro Präkonditionierung von MSC vielversprechend sein. Die Verwendung von Niclosamid hingegen sollte vermieden werden, die Wirkung von Gedunin müsste genauer untersucht werden.
This short paper gives a brief overview of the manifestly covariant canonical gauge gravity (CCGG) that is rooted in the De Donder-Weyl Hamiltonian formulation of relativistic field theories, and the proven methodology of the canonical transformation theory. That framework derives, from a few basic physical and mathematical assumptions, equations describing generic matter and gravity dynamics with the spin connection emerging as a Yang Mills-type gauge field. While the interaction of any matter field with spacetime is fixed just by the transformation property of that field, a concrete gravity ansatz is introduced by the choice of the free (kinetic) gravity Hamiltonian. The key elements of this approach are discussed and its implications for particle dynamics and cosmology are presented. New insights: Anomalous Pauli coupling of spinors to curvature and torsion of spacetime, spacetime with (A)dS ground state, inertia, torsion and geometrical vacuum energy, Zero-energy balance of the Universe leading to a vanishing cosmological constant and torsional dark energy.
Exploring strategies to improve the reverse beta-oxidation pathway in Saccharomyces cerevisiae

Microbes are the most diverse living organisms on Earth, with various metabolic adaptations that allow them to live in different conditions and produce compounds with different chemical complexity. Microbial biotechnology exploits the metabolic diversity of microorganisms to manufacture products for different industries. Today, the chemical industry is a significant energy consumer and carbon dioxide emitter, with processes that harm natural ecosystems, like the extraction of medium-chain fatty acids (MCFAs). MCFAs are used as precursors for biofuels, volatile esters, surfactants, or polymers in materials with enhanced properties.
However, their current extraction process uses large, non-sustainable monocultures of coconut and palm trees. Therefore, the microbial production of MCFAs can help reduce the current environmental impact of obtaining these products and their derivatives.
In nature, fatty acids are mostly produced via fatty acid biosynthesis (FAB). However, the reverse β-oxidation (rBOX) is a more energy-efficient pathway compared to FAB. The rBOX pathway consists of four reactions, which result in the elongation of an acyl-CoA molecule by two carbon units from acetyl-CoA in each cycle. In this work we used Saccharomyces cerevisiae, an organism with a high tolerance towards toxic compounds, as the expression host of the rBOX pathway to produce MCFAs and medium-chain fatty alcohols (MCFOHs).
In the first part of this work, we expanded the length of the products from expressing the rBOX in the cytosol and increased the MCFAs titres. First, we deleted the major glycerol-3-phosphate dehydrogenase (GPD2). This resulted in a platform strain with significantly reduced glycerol fermentation and increased rBOX pathway activity, probably due to an increased availability of NADH. Then, we tested different combinations of rBOX enzymes to increase the length and titres of MCFA. Expressing the thiolase CnbktB and β-hydroxyacyl-CoA dehydrogenase CnpaaH1 from Cupriavidus necator, Cacrt from Clostridium acetobutylicum and the trans-enoyl-CoA reductase Tdter (Treponema denticola) resulted in hexanoic acid as the main product.
Expressing Cncrt2 (C. necator) or YlECH (Y. lipolytica) as enoyl-CoA hydratases resulted in octanoic acid as the main product. Then, we integrated the octanoic (Cncrt2 or YlECH) and the hexanoic acid (Cacrt)-producing variants in the genome of the platform strain and we achieved titers of ≈75 mg/L (hexanoic acid) and ≈ 60 mg/L (octanoic acid) when growing these strains in a complex, highly buffered medium. These are the highest titers of octanoic and hexanoic acid obtained in S. cerevisiae with the rBOX. Additionally, we deleted TES1 and FAA2 to prevent competition for butyryl-CoA and degradation of the produced fatty acids, respectively.
However, these deletions did not improve MCFA titers. In addition, we tested two dual acyl-CoA reductase/alcohol dehydrogenases (ACR/ADH), CaadhE2 from C. acetobutylicum and the putative ACR/ADH EceutE from Escherichia coli, in an octanoyl-CoA-producing strain to produce MCFOH. As a result, we produced 1-hexanol and 1-octanol for the first time in S. cerevisiae with these two enzymes. Nonetheless, the titres were low (<10 mg/L and <2 mg/L, respectively), and four-carbon 1-butanol was the main product in both cases (>80 mg/L). This showed the preference of these two enzymes for butyryl-CoA.
In the second part of this work, we expressed the rBOX in the mitochondria of S. cerevisiae to benefit from the high levels of acetyl-CoA and the reducing environment in that organelle. First, in an adh-deficient strain, we mutated MTH1, a transcription factor regulating the expression of hexose transporters, and deleted GPD2. This resulted in a strain with a reduced Crabtree effect and, therefore, an increased carbon flux to the mitochondria. We partially validated the increased flux to the mitochondria by expressing the ethanol-acetyltransferase EAT1 from Kluyveromyces marxianus in this organelle. This resulted in a higher isoamyl acetate production in the MTH1-mutant strain. Isoamyl acetate is synthesised by Eat1 from acetyl-CoA and isoamyl alcohol, a product of the metabolism of amino acids in the mitochondria. Then, we targeted different butyryl-CoA-producing rBOX variants to the mitochondria, and we used the production of 1-butanol and butyric acid as a proof-of-concept. The strong expression of all the enzymes was toxic for the cell, and the highest butyric acid titres (≈ 50 mg/L) in the mitochondria from the rBOX were obtained from the weak expression of the pathway. The highest 1-butanol titers (≈ 5 mg/L) were obtained with the downregulation of the mitochondrial NADH-oxidase NDI1. However, this downregulation led to a non-desirable petite phenotype.
In summary, we produced hexanoic and octanoic acid for the first time in S. cerevisiae using the rBOX and achieved the highest reported titers of hexanoic and octanoic acid so far using this pathway in S. cerevisiae. In addition, we successfully compartmentalised the rBOX in the mitochondria. However, competing reactions, some of them essential for the viability of the cell, limit the use of this organelle for the rBOX.
Die Appendizitis stellt mit einer Inzidenz von 115 pro 100000 Einwohnern in Deutschland eine der häufigsten Ursachen für ein akutes Abdomen dar. Bakterielle Infektionen sind ein wesentlicher Faktor für die postoperative Morbidität nach Appendektomie.
Ziel dieser prospektiven Studie war es, das Keimspektrum und insbesondere die Prävalenz resistenter Keime bei der akuten Appendizitis zu bestimmen und die Auswirkungen resistenter Keime auf das Auftreten infektiöser Komplikationen zu analysieren. Alle erwachsenen Patient*innen mit akuter Appendizitis, die zwischen April 2022 und Juli 2023 am Universitätsklinikum Frankfurt operativ behandelt wurden, wurden prospektiv eingeschlossen. Das Keimspektrum der Appendix und die Häufigkeit von MRE in Rektalabstrichen wurden analysiert. Die klinischen Daten wurden extrahiert, um die Korrelation des Keimspektrums mit dem Auftreten von postoperativen Komplikationen, Dauer und Art der Antibiotikatherapie und dem postoperativen Verlauf zu evaluieren. 30 Tage nach der Operation wurde ein Follow-up durchgeführt. Insgesamt wurden 105 Patient*innen in die Studie eingeschlossen.
In den Appendixabstrichen gelang ein Erregernachweis bei 67,6 % aller Fälle. Hierbei betrug die Prävalenz von Keimen mit Antibiotikaresistenz 43,8 %, Multipler-Antibiotikaresistenz (MAR) 27,6 % und bei 5,7 % der Fälle gelang ein MRE-Nachweis nach CDC im Appendixabstrich. Beim MRE-Screening konnten im Rektalabstrich bei 11,4 % der Patient*innen ein MRE nachgewiesen werden. In vier Fällen zeigte sich eine Übereinstimmung zwischen rektalen und appendikulären Abstrich, somit betrug die Sensitivität des MRE-Screenings für einen Nachweis multiresistenter Keime in der Appendix lediglich 33,3 % bei einer Spezifität von 86,7 %. In einer univariaten Analyse konnte das Vorliegen eines Diabetes mellitus als Risikofaktor für das Auftreten von Keimen mit Cefuroxim- Resistenz, Multipler-Antibiotikaresistenz (MAR) und MRE identifiziert werden.
Insgesamt erhielten 47,6 % aller Patient*innen postoperativ eine Antibiotikatherapie, von denen erfolgte bei 46 % eine Umstellung der empirischen Antibiose auf eine antibiogramm-gerechte Therapie nach Erhalt des mikrobiologischen Befundes. Insbesondere Patient*innen mit komplizierter Appendizitis erhielten postoperativ eine Antibiotikatherapie, wobei 28 % eine Antibiotikaeskalation benötigten.
Patient*innen, deren Appendixabstriche eine Resistenz gegen maximal ein Antibiotikum aufwiesen, wurden als nicht multiple Resistenz (Nicht-MAR) definiert und mit Patient*innen verglichen, deren Keime mit multipler Antibiotikaresistenz (MAR) waren. Die MAR-Gruppe zeigte im Vergleich zur Nicht-MAR-Gruppe eine höhere Inzidenz postoperativer Komplikationen, insbesondere eine erhöhte Inzidenz von Clavien-Dindo Grad 3 Komplikationen sowie von tiefen postoperativen Wundinfektionen (CDC Grad A3). Weiterhin benötigten Patient*innen der MAR-Gruppe häufiger eine postoperative Antibiotikatherapie und es erfolgte häufiger eine Eskalation der antibiotischen Therapie. Dies ging auch mit einem signifikant verlängerten Krankenhausaufenthalt einher.
Zusammenfassend kann festgestellt werden, dass eine bakterielle Infektion mit Multipler Antibiotikaresistenz bei der akuten Appendizitis häufig auftritt und die Morbidität nach Appendektomie beeinflusst. Die mikrobiologische Untersuchung mittels Appendixabstrich bei operativ behandelten Fällen von akuter komplizierter Appendizitis, die eine postoperative Antibiotikatherapie erfordern, könnte somit eine gezielte und kürzere Antibiotikatherapie ermöglichen. Dies könnte dazu beitragen, längere Krankenhausaufenthalte zu vermeiden, den unnötigen Einsatz unwirksamer Antibiotika zu reduzieren und die Kosten im Gesundheitswesen zu senken.
Hidradenitis suppurativa ist eine multifaktoriell-bedingte chronisch entzündliche Hauterkrankung, die durch eine Okklusion der Talgdrüseneinheit des Haarfollikels entsteht. Aus der anschließend mit Entzündung einhergehenden Ruptur des Haarfollikels entwickeln sich entzündliche Knoten, Abszesse und Fistelgänge. (39–41) In der weiteren Progression der Erkrankung kommt es zur Störung der Hautarchitektur und fibrotischen Narbenbildungen. (52) Durch Untersuchungen des entzündlichen Infiltrates konnte bereits die Beteiligung einer Reihe von Immunzellen und Entzündungsmediatoren identifiziert werden. Hierzu zählen Makrophagen, neutrophile Granulozyten, Dendritische Zellen, Lymphozyten, IL-1β sowie TNF-α. (41,45,46,51,53) Da die genaue Pathophysiologie der Hidradenitis suppurativa bislang unzureichend aufgeklärt ist, gibt es aktuell keine kausale Therapiemöglichkeit für die Betroffenen. (52) Die Wahl der Therapie wird anhand der Bewertung des Schweregrades nach Hurley getroffen. (25) Obwohl meisten Patientinnen und Patienten von der milden bis mittelschweren Form der Hidradenitis suppurativa (Hurley I und Hurley II) betroffen sind (98), werden die meisten Arzneimittel für die Behandlung von Hurley II und Hurley III von den Leitlinien empfohlen. Zu den empfohlenen Medikamenten gehören u. a. Rifampicin, meist in Kombination mit Clindamycin, sowie der TNF-α-Inhibitor Adalimumab (18,85), welche effektiv und systemisch wirken. Durch die entstehenden Nebenwirkungen wäre die Behandlung der milden bis mittelschweren Hidradenitis suppurativa mit diesen Medikamenten allerdings unverhältnismäßig. Um jedoch den im Verlauf der Krankheit entstehenden Hautdestruktionen vorbeugen zu können, müssten die Medikamente möglichst früh eingesetzt werden. (228) Vor diesem Hintergrund sollte in Kooperation mit dem Institut für Pharmazeutische Technologie der Goethe Universität Frankfurt eine Rifampicin-Nanoformulierung zur Behandlung milder bis mittelschwerer Hidradenitis suppurativa entwickelt und im Labor der dermatologischen Klinik präklinisch validiert werden.
Daher wurde im ersten Teil der vorliegenden Arbeit ein Epidermismodell aus der Haut betroffener Patientinnen und Patienten generiert, um die Rifampicin-Nanoformulierung validieren zu können. Dieses wies eine mehrschichtige Epidermis mit allen wichtigen Differenzierungsmarkern ähnlich der läsionalen Hidradenitis suppurativa auf und schüttete die proinflammatorischen Zytokine IL-1β und TNF-α aus. Als weiteres Modell wurden ex vivo Explantate aus läsionaler Hidradenitis suppurativa Haut etabliert. Für die Bewertung der Validität der Explantatkulturen wurde die Morphologie und Integrität der Epidermis mittels Hämatoxylin-Eosin sowie der Proliferationsmarker Ki-67 näher beleuchtet. Im zweiten Teil dieser Arbeit wurden Untersuchungen für die Festlegung eines Konzentrationsbereichs, unter Verwendung von Rifampicin in DMSO gelöst als in vitro Behandlung, durchgeführt.
Hierbei wurde gezeigt, dass die eingesetzten Konzentrationen keine negativen Effekte bezüglich Proliferationsfähigkeit der Keratinozyten oder Apoptoseinduktion ausüben. Die Behandlung von nicht entzündlichen Epidermismodellen sowie Explantatkulturen mit der Rifampicin-Nanoformulierung mit einem Wirkstoffgehalt von 0,3 % führte ebenfalls zu keinen Proliferationsverlusten, induzierte keine Apoptose oder Zytotoxizität und hatte keinen Einfluss auf die Differenzierung der Keratinozyten. Im letzten Teil der Arbeit sollte die Wirksamkeit der Rifampicin-Nanoformulierung näher beleuchtet werden. Als Antibiotikum inhibiert Rifampicin die DNA-abhängige RNA-Polymerase von Bakterien (107,108), weshalb es bei der Behandlung der Tuberkulose eingesetzt wird. (111–113) Die Hidradenitis suppurativa ist aber primär keine Infektionskrankheit, sondern eine von Bakterien getriggerte, entzündliche Erkrankung. (19,52,82) Aus diesem Grund ist der positive Effekt der systemischen Rifampicintherapie vermutlich vielmehr auf eine antiphlogistische Wirkung zurückzuführen. Diese Überlegung wird durch mehrere Artikel gestützt, die zeigten, dass Rifampicin die Ausschüttung von IL-1β und TNF-α in unterschiedlichen in vitro und in vivo Modellen hemmt. (89–91,96,97) Die anschließenden Untersuchungen zur Wirksamkeit einer in vitro Rifampicinbehandlung bestätigten die antientzündliche Wirkung in den Explantatkulturen indem es die Sekretion von IL-1β, IL-6, IL-8, IL-10 und TNF-α verminderte. Ebenso senkte die Rifampicin-Nanoformulierung die Ausschüttung von IL-1β in den ex vivo Explantaten, was somit die in der klinischen Praxis beobachteten antiinflammatorischen Wirkung von Rifampicin belegt. Des Weiteren stellte sich in Untersuchungen heraus, dass Rifampicin zu einer reduzierten Zahl CD4(+)-T-Zellen führte, aber auf die CD3(+)-T-Zellen keine Auswirkungen hatte, was auf eine Veränderung des T-Zell Phänotyps hinweist.
Aufgrund der vorliegenden Ergebnisse zur Validierung der Rifampicin-Nanoformulierung, spricht nichts gegen die Testung der Rifampicin-Nanoformulierung in einem individuellen Heilversuch am Menschen.
We report results on an elastic cross section measurement in proton–proton collisions at a center-of-mass energy √𝑠 = 510 GeV, obtained with the Roman Pot setup of the STAR experiment at the Relativistic Heavy Ion Collider (RHIC). The elastic differential cross section is measured in the four-momentum transfer squared range 0.23 ≤ −𝑡 ≤ 0.67 GeV2. This is the only measurement of the proton-proton elastic cross section in this 𝑡 range for collision energies above the Intersecting Storage Rings (ISR) and below the Large Hadron Collider (LHC) colliders. We find that a constant slope 𝐵 does not fit the data in the aforementioned 𝑡 range, and we obtain a much better fit using a second-order polynomial for 𝐵(𝑡). This is the first measurement below the LHC energies for which the non-constant behavior 𝐵(𝑡) is observed. The 𝑡 dependence of 𝐵 is also determined using six subintervals of 𝑡 in the STAR measured 𝑡 range, and is in good agreement with the phenomenological models. The measured elastic differential cross section d𝜎∕dt agrees well with the results obtained at √𝑠 = 540 GeV for proton–antiproton collisions by the UA4 experiment. We also determine that the integrated elastic cross section within the STAR 𝑡-range is 𝜎f id el = 462.1 ± 0.9(stat.) ± 1.1(syst.) ± 11.6(scale) 𝜇b.
The differential cross section for 𝑍0 production, measured as a function of the boson’s transverse momentum (𝑝T), provides important constraints on the evolution of the transverse momentum dependent parton distribution functions (TMDs). The transverse single spin asymmetry (TSSA) of the 𝑍0 is sensitive to one of the polarized TMDs, the Sivers function, which is predicted to have the opposite sign in 𝑝 + 𝑝 → 𝑊 ∕𝑍 + 𝑋 from that which enters in semi-inclusive deep inelastic scattering. In this Letter, the STAR Collaboration reports the first measurement of the 𝑍0∕𝛾∗ differential cross section as a function of its 𝑝T in 𝑝+𝑝 collisions at a center-of-mass energy of 510 GeV, together with the 𝑍0∕𝛾∗ total cross section. We also report the measurement of 𝑍0∕𝛾∗ TSSA in transversely polarized 𝑝+𝑝 collisions at 510 GeV.
Although, during the past decades, substantial advances emerged in identifying major local and systemic factors contributing to initiation and progression of osteoarthritis (OA), some neuroendocrine mechanisms are still not understood or even neglected when thinking about novel therapeutic options. One of which is the sympathetic nervous system that exhibits various OA-promoting effects in different tissues of the joint. Interestingly, the β2-adrenoceptor (AR) mediates the majority of these effects as demonstrated by several in vitro, in vivo as well as in clinical studies. This review article does not only summarize studies of the past two decades demonstrating that the β2-AR plays an OA-promoting role in different tissues of the joint but also aims to encourage the reader to think about next-level research to discover novel and innovative preventive and/or therapeutic strategies targeting the β2-AR in OA.

Polygene Risikoscores (PRS) integrieren zahlreiche Einzelnukleotid-Polymorphismen (SNP) von meist geringer Effektstärke, um Auskunft über das Erkrankungsrisiko bestimmter Krankheiten zu geben. In dieser Arbeit wurde der PRS zur genetisch generalisierten Epilepsie (GGE) von Leu et al. aus dem Jahr 2019 untersucht, um festzustellen, ob über das Erkrankungsrisiko hinaus noch Korrelationen mit weiteren phänotypischen Eigenschaften von Patienten bestehen. Der Nachweis solcher Zusammenhänge würde eine Prädiktionsfähigkeit des GGE-PRS demonstrieren, die perspektivisch ein Potential für dessen klinische Anwendbarkeit, beispielsweise im Sinne der personalisierten Medizin, aufzeigen könnte.
Die Identifizierung neuer Korrelationen sollte durch Vergleich der Phänotypen von zwei Gruppen von GGE-Patienten mit extrem hohen, beziehungsweise extrem niedrigen PRS-Werten erfolgen. Hierfür wurden von 2256 Patienten aus der Datenbank von Epi25, einem internationalen Forschungskollaborativ zur Erforschung der Relevanz genetischer Faktoren bei der Entwicklung von Epilepsie, die Patienten mit den höchsten (n=59) und den niedrigsten (n=49) GGE-PRS-Werten ausgewählt. Für diese 108 Patienten wurden retrospektive klinische Daten von den jeweiligen Behandlungszentren akquiriert. Hierzu wurde den Studienleitern der Zentren ein Questionnaire mit Fragen zu zahlreichen phänotypischen Parametern der Patienten übermittelt. Die Rücklaufrate war mit 54% gut.
Die so eingeholten Patientendaten wurden anschließend mittels Exaktem Test nach Fisher und Wilcoxon-Rangsummentest statistisch analysiert, um Unterschiede zwischen den Phänotypen beider Gruppen nachzuweisen. Im Falle der Pharmakoresistenz zeichneten sich hierbei zunächst signifikante Unterschiede ab, die ein selteneres Auftreten dieser Eigenschaft für Patienten mit hohen GGE-PRS-Werten implizierten. Diese Ergebnisse waren jedoch nach einer Bonferroni-Korrektur und bei Validierung in einer größeren Kohorte (n=825) nicht mehr signifikant. Für die anderen untersuchten Parameter waren ebenfalls keine signifikanten Unterschiede nachweisbar.
Das Ergebnis, dass für keinen der untersuchten Parameter signifikante Differenzen bestanden, obwohl zwei Kohorten mit extrem gegensätzlichen PRS-Werten untersucht wurden, spricht gegen eine Verwendung des aktuell verfügbaren GGE-PRS als prädiktiver Biomarker über das Erkrankungsrisiko hinaus und somit gegen dessen klinische Anwendbarkeit. Jedoch können die nicht-signifikanten Korrelationen im Falle der Pharmakoresistenz als Hinweis verstanden werden, dass im Bereich der Pharmakotherapie Zusammenhänge zwischen Score und Phänotyp bestehen könnten, die weiterer Untersuchungen in zukünftigen Studien bedürfen. Bei Verwendung eines verbesserten GGE-PRS mit zusätzlichen risikoassoziierten SNP und verfeinerter Wichtung der Effektstärken sowie größerer Kohorten könnten in diesem Bereich möglicherweise auch signifikante Zusammenhänge nachweisbar werden.
How can older adults participate equally in digitisation processes across Europe, and what inclusive research strategies are needed? This Zine summarizes findings from a “Research Innovation Lab on Ageing in a Digital Age”, funded by the VolkswagenStiftung, aiming to bring together 29 docs and postdocs anchored in 26 different disciplines coming from 11 countries, at all stages of their work, to address cutting edge questions relating to ageing in a digital age. Five groups worked together over five days in Frankfurt, Germany, in July 2023 in a creative and interactive hackathon, specific to developing non-technical solutions to social issues of this topic. Moreover, four distinguished experts presented keynote speeches and proposals from various conceptual, methodological and empirical perspectives.
L'obiettivo del presente articolo è quello di offrire una visione complementare a quella abituale del romanzo "Die Entdeckung der Currywurst" di Uwe Timm in cui vengano privilegiati gli aspetti del romanzo che lo identificano come opera di World Literature. Se fino ad ora il romanzo è stato principalmente letto come un romanzo tedesco di guerra, con questa lettura si intende far emergere gli aspetti che di esso oltrepassano i confini nazionali e linguistici. Per fare questo si osserveranno in breve i punti di contatto fra la WL e la letteratura postcoloniale, dalla quale la WL stessa ha origine. In particolare si concentrerà l'attenzione sugli elementi del romanzo che evidenziano il rapporto con le letterature e culture di oltre confine, come la cornice narrativa, in cui la narrazione principale si iscrive; le contaminazioni letterarie che emergono dal racconto e infine i numerosi esempi di relazione fra centro e periferia. Inoltre, verrà evidenziato come il periodo storico in cui il romanzo è ambientato, ovvero quello della 'Stunde Null', abbia esso stesso dei contatti intrinseci con le teorie postcoloniali. Si mostrerà come la tesi dell'afferenza del presente romanzo alla WL, sia sostenuta dal fatto che anche attraverso romanzi precedenti, Uwe Timm mostra un interesse spiccato nei confronti delle realtà degli altri paesi e continenti e dell’eredità coloniale dell'Europa.
E.T.A. Hoffmanns Erzählung "Der Sandmann" ist dank Freuds Interpretation zu einem seiner wichtigsten Werke geworden. Die Novelle, die die Leser mit dem jungen Studenten Nathanael, der mit sich ein Kindheitstrauma trägt, bekannt macht, ist ein komplexes Werk, das auch den weltbekannten Mediziner fasziniert hat. Diese Arbeit befasst sich mit der schon erwähnten Novelle, mit Freuds Artikel "Das Unheimliche" und mit der sehr relevanten Verfilmung aus dem Jahr 2012. Das Motiv des Unheimlichen kommt in dieser Arbeit als eine Kontaktstelle vor, die auf drei verschiedenen Ebenen interpretiert wird: aus der Perspektive des Unheimlichen in der Novelle, aus Sigmund Freuds Perspektive und wie es im Film auftaucht. Ziel dieser Arbeit ist es zu beweisen, dass das Motiv des Unheimlichen in der Verfilmung anders als im Buch begriffen werden kann, d.h. dass Freuds Motiv des Unheimlichen im Text als gruseliger und verwirrender empfunden werden kann.

Arbeit und Spiel fungieren im allgemeinen Sprachgebrauch als Gegensätze. Spielen wird im Kontext dieser Oppositionsbeziehung als autotelische Handlung, also als eine freiwillige Beschäftigung mit unproduktivem Charakter, definiert, die primär der Freude oder Entspannung dient. Arbeiten hingegen wird als zweckorientierte Tätigkeit gefasst, die mit Ernsthaftigkeit und Mühsal assoziiert wird. In einer sich verändernden Arbeitswelt lösen sich diese binären Zuordnungen, sofern sie in dieser Schärfe überhaupt jemals bestanden haben, jedoch zunehmend auf. Arbeit und Spiel verschränken sich, sie bilden Hybridformen aus, bei denen sich Verschiebungen von Spielenden zu Arbeitenden und umgekehrt beobachten lassen. Die Etablierung der Spieleindustrie als Wirtschaftsbranche oder die Professionalisierung des Wettkampfsports bringen beispielsweise Formen des Play-as-Work hervor: "Sämtliche Tätigkeiten mit einer unterstellten Wirkabsicht können demnach als Arbeit begriffen werden. Daher können auch aus traditionellen Gründen oder aus Unterhaltungs- und Zerstreuungsgründen ausgeführte Tätigkeiten nunmehr als Arbeit verstanden werden, wenn ihnen eine Wirkabsicht unterstellt werden kann." Zudem werden in Arbeitsbereichen der postfordistischen Wissensökonomie ludische Elemente wie Kreativität, Improvisation oder Risikobereitschaft in einer Abwendung von der normierten Rationalisierung von Arbeit des Taylorismus aufgewertet und spielhaftes Arbeiten ohne kodifizierte Regeln (wieder) als Mittel wirtschaftlicher Produktivität erkannt. Seit den 1990er Jahren lassen sich ferner Tendenzen einer zunehmenden Entgrenzung und Subjektivierung von Erwerbstätigkeit ausmachen; hierbei wird die mit der Industrialisierung entstandene Trennung von Arbeit und Leben abgeschwächt und das Konzept Arbeit neu definiert, denn diese wird nun nicht mehr vornehmlich negativ als Plage oder göttliche Strafe gerahmt, sondern positiv als Mittel der Selbstverwirklichung konnotiert. Arbeit soll infolgedessen spielend von der Hand gehen und Vergnügen bereiten. Auf diese Weise wird eine work-as-play-Mentalität erzeugt, die die Differenz von Ernst und Spiel in der Betrachtungsweise der Handlung festmacht: Es hängt vom Ausführenden ab, ob eine Tätigkeit als Spiel oder als Arbeit klassifiziert wird. So kann letztlich jede Aktivität einfach zum Spiel erklärt werden. [...] Im Fokus dieses Artikels soll jedoch eine andere Form der Hybridisierung von Arbeit und Spiel stehen: die Repräsentation von Büroarbeit im Computerspiel. [...] Untersucht werden soll im Folgenden, welche Inszenierungsstrategien und Spielmechaniken eingesetzt werden, um das Büro spielbar zu machen. Wie wird das Setting für das Gameplay funktionalisiert und wie werden Arbeitsprozesse ludifiziert? Zu welcher Form des Spiels lädt das Büro ein? Der Begriff der Verblendung, ein oft implizit immer wiederkehrendes Thema in der Literatur, Philosophie, und auch in der Religion, hat eine komplexe Bedeutungsebene, die es vielleicht wohl zu bewahren gilt, anstatt sie zu fixieren und damit die Lesemöglichkeiten, die er eröffnen kann, zu reduzieren. Das Wort, das Offenes (etwa eine Hingabe an einen Glauben, die auch eine Selbsttäuschung, ein Fehler sein kann, aber auch Mut bedeuten kann) und Verborgenes (eine mehr oder weniger bewusste Täuschung anderer) zugleich andeutet, entzieht sich durch diese doppelte Struktur von 'Oberfläche' und 'Hintergrund' in gewisser Weise, ohnehin einer festen BeDeutungsgebung. In besonderer Weise bleibt der Begriff mit dem als 'Verliebtheit' umschriebenen Zustand als einer ambivalenten un/wahren, irrational und affektiv besetzten, relationalen, inneren Vorstellung und äußeren Projektion des Selbst und eines Anderen verbunden. Der Begriff kann die Hingabe an eine (trügerische) Romantik oder eine (trügerische) Hingabe an eine Romantik meinen, der ein Selbst und ein Anderes in eine affektive Beziehung zueinander setzt, und so auch die Möglichkeit des Unheimlichen, das Hineinbrechen eines Trug(-Bild)-es kennzeichnen. Dies wiederum ist engstens mit Angst und Wut, den oft als destruktiv dargestellten Affekten und affektiv besetzten Effekten verbunden. Verblendung ist somit auch ein Begriff, der Vorstellungen von 'Rationalität' und (affektiv-besetzter) 'Irrationalität', von 'Richtigkeit' und 'Falschheit' beinhaltet.
This paper relies on an unedited and unpublished nineteenth century love correspondence of a heterosexual couple from the German speaking area. The aim of this study is to contribute to the knowledge regarding the nineteenth century love experience of ordinary and unknown lovers. In fact, while there are plenty of books on love correspondences of famous personalities, little research has been dedicated to love letters and romantic experiences of 'ordinary and unknown' people. For this reason the main aim of this article is to shed light on love stories and love experiences that otherwise will fall into the abyss of oblivion. A new theory regarding the love experience in the nineteenth will be proposed: in this century love was perceived more in its material than in its abstract nature; I argue that in the nineteenth love was more about what people did, than to what they said. Lovers are in constant need of material and 'seeable' proof in order to perceive the love of their partners as real and authentic. The examples extrapolated from the following correspondence will corroborate this statement. Furthermore, this article has the purpose to underline the great value of love letters not only from an historical perspective (being evidence of past lives and dynamics), but also and more importantly from a cultural and societal one: analyzing love letters means to acquire knowledge not only about cultural and societal dynamics, but also and more importantly to add knowledge to the love discourse. In fact, they say a lot about the way people talked, expressed and materialized love in their daily lives. Correspondences without any publication in view are the most precious ones because they represent an unregulated and more spontaneous expression 'of the language of the heart'.
Kriminalität und deren Bekämpfung unterliegen seit jeher permanenten Veränderungen. Durch den gesellschaftlichen Wandel und die damit einhergehende Weiterentwicklung von technischen Möglichkeiten werden fortlaufend neue Modi Operandi ermöglicht, auf welche die Polizei mit teilweise ebenfalls neuen kriminalistischen Mitteln reagiert. Entwicklungen in der Kriminalität und der Kriminalistik können somit als sich gegenseitig beeinflussende Prozesse angesehen werden. Allerdings findet dieser Wandel nicht kontinuierlich statt, sondern tritt in Schüben auf, die in der Regel auf besonders kriminalitätsrelevante Ereignisse zurückzuführen sind. In diesem Zusammenhang verändert sich die Polizei, sowohl in ihrer Organisation als auch in ihren Analysen zur Entwicklung der Kriminalität und in ihren Methoden zu deren Bekämpfung, teilweise grundlegend.
In einem ersten, konzeptionellen Teil wird anhand von ausgewählten Kriminalitätsformen und den damit verbundenen Ermittlungspraktiken ein Überblick der gegenseitigen Beeinflussung von Kriminalität und Kriminalistik gegeben. Darauf aufbauend wird im zweiten Teil der aktuelle Stand dieses 'Evolutionsprozesses' anhand der Implementierung von Predictive-Policing-Systemen in die Polizeiarbeit diskutiert. Denn darin zeigt sich nahezu idealtypisch die gegenseitige Beeinflussung von Kriminalität und Kriminalistik wie auch der digitale Wandel und die damit verbundenen Gefahren. Polizeiarbeit scheint sich im Ergebnis dieser Veränderungsprozesse von einer ursprünglich eher auf den Einzelfall bezogenen, subjektiven Sichtweise auf Kriminalität und deren Verfolgung (zum Beispiel durch kriminalistische Taktik und List in der Vernehmung) auf eine nunmehr eher allgemeine, abstraktere Betrachtung zu verschieben ('Abstract Police').
Inwiefern schließen Open Source Investigations eine Revision des Indizienparadigmas ein? Der Aufsatz beantwortet diese Frage, indem er anhand von Fallbeispielen von Rechercheagenturen (bellingcat und Forensic Architecture) und investigativen Fotografen (Trevor Paglen und Edmund Clark) eine Verschiebung im Spurbegriff herausarbeitet: Im Rahmen digitaler Recherchen sind Spuren durch Datenverarbeitung hervorgebrachte mediale Artefakte, sie werden also nicht am Tatort vorgefunden, sondern rechnerisch produziert. In den Falldarstellungen und Methodenreflexionen der Akteur:innen können dabei verschiedene Modi der virtuellen Spur unterschieden werden.
Der Beitrag thematisiert die literarische Darstellung virtueller Investigationen, welche eine performativ-schauspielerische Wiederholung von Tatgeschehnissen beinhalten. Er analysiert insbesondere die Rolle von Zufallsmotiven und -aspekten in solchen Darstellungen und deren metapoetische Bedeutung im Rahmen einer Gattungspoetik des Kriminalromans. Als Fallbeispiele dienen hierfür Friedrich Dürrenmatts 'Requiem auf den Kriminalroman' "Das Versprechen" und Stanisław Lems Roman "Der Schnupfen". Im Vergleich der beiden Texte zeigt sich unter anderem, dass die Idee eines genauen 're-enactment' von Tathergängen auf der Grundlage virtueller Rekonstruktionen in beiden Romanplots nicht direkt zum gewünschten Ergebnis führt, dass aber in Lems Roman eine Integration des Faktors Zufall in virtuelle Ermittlungsprozesse und in die Poetik des Kriminalromans insgesamt sehr viel positiver gewertet wird, während der Einfluss des Zufalls bei Dürrenmatt ebenfalls als unhintergehbar, aber nicht als uneingeschränkt begrüßenswert perspektiviert wird.
Lange hat sich die Feststellung von Subjekten auf den menschlichen Körper gerichtet, auf dessen Spur, besonderes Merkmal, Fingerabdruck oder DNA-Profil. Derweil verliert sich dieser indexikalische Zusammenhang in digitalen Erkennungsverfahren, einerseits indem sie nicht länger ein Indiz auf einen unverwechselbaren Körper beziehen, sondern dessen Bild nunmehr mit den Pixel-Verteilungen in tausenden anderen Bilddateien abgleichen, andererseits weil sie, noch vor der Identifizierung einer Person, überhaupt erst einen "Körper" als solchen registrieren müssen. Damit aber verschieben sich all jene subversiven Praktiken, die den Menschen der Detektion zu entziehen trachten: Statt bloßer Maskierung gilt es jetzt, den Körper selbst zu tilgen - durch technische Verwischungen, Oberflächenmodulationen oder Bewegungsmuster außerhalb der KI-Trainingsraster. Derlei Strategien des Entzugs werden in der zeitgenössischen Videokunst erprobt, etwa in Hito Steyerls "How Not to Be Seen" oder Liam Youngs "Choreographic Camouflage". Damit stellt sich zudem die grundsätzliche Frage nach der Verfügbarkeit und Neuform(at)ierung des Körpers im Digitalen; hier nämlich verwandelt er sich vom Zeichen- und Faktenreservoir in Virtualität und stetes Werden, die ihr politisches Potential auch weit jenseits aller Überwachungstechnologie entfalten.
Der Beitrag konzentriert sich auf einen blinden Fleck in Carlo Ginzburgs Indizienparadigma. Ginzburg beschränkt die Ermittlung von Wissen im Bereich des Rechts auf die Erhebung materieller Indizien der Vergangenheit. Im größeren Zusammenhang der staatlichen Rationalität des 18. Jahrhunderts wird jedoch deutlich, dass bei Ginzburg die präventive und damit zukunftsgewandte Komponente der Guten Policey fehlt. Um potentielle Gefahren abzuwehren und bestenfalls unmöglich zu machen, werden im Dienste der Generalprävention Spuren einer möglichen Zukunft lesbar gemacht. Doch führt die bloße Virtualität einer potentiellen Zukunft zu einer Universalisierung des Verdachts und alle Menschen werden zu potentiellen Verbrechern. Mit Schiller und Fichte macht der Beitrag daher auch die Grenzen staatlicher Fürsorge namhaft. Im Bereich der Literaturwissenschaft entspricht den Investigationen der Guten Policey die rezeptionsästhetische Theorie. Sie ermittelt das Bedeutungspotential, das einem Werk immer schon innewohnt, aber erst in der späteren Rezeption entfaltet wird. Der Bedrohung einer Universalisierung des Verdachts entspricht bei der Rezeptionsästhetik die Gefahr, das Bedeutungspotential zu entgrenzen, womit die Lesart der Indizien beliebig würde.
Das ludische Dispositiv virtueller Investigationen : auf Spurensuchen in interaktiven Krimi-Formaten

In interaktiven Kriminalerzählungen verknüpfen sich Medialität und Ästhetik, wobei Computer als Werkzeuge der Ermittlungsarbeit neue Perspektiven auf polizeiliche Effizienz und agency eröffnen. Narrative Spiele nutzen Technologie und Medien häufig auf innovative und selbstreflexive Weise, um immersive Spielwelten und ludische Herausforderungen für die Spielenden zu schaffen. Dieser Artikel verfolgt die historische Entwicklung von Kriminalspielen anhand einzelner Beispiele. Denn diese weisen bedingt durch ihr Erscheinungsdatum spezifische Formen technischer und ästhetischer Verfahren auf, werfen unterschiedliche Fragen zur Natur von Wissen, Medien und Wahrheit auf und eröffnen unterschiedliche Perspektiven auf die Wechselwirkungen zwischen virtueller und realer Kriminalität sowie auf mögliche Einflüsse auf reale kriminologische Praktiken.
Wenn im Rahmen von investigativen Recherchen auf öffentlich und digital verfügbare Daten zurückgegriffen wird, spielen Bilder der physischen Tatumgebung häufig eine wesentliche Rolle. Ins Internet gestellte Fotos und Videos von Ereignissen im öffentlichen Raum oder topographische Luftbilder im Vorher-Nachher-Vergleich gelten dabei oft als beste Beglaubigung eines Geschehens. Der technische Charakter und die Georeferenzierung verleihen den Bildern eine so hohe Überzeugungskraft, dass sie als Grundlage für rekonstruierende 3D-Modelle und Simulationen verwendet werden - auch wenn ihre Glaubwürdigkeit in den letzten Jahren durch die Möglichkeit der Bildmanipulation oder der gezielten Fehlinterpretation stark gelitten hat. Anhand von Fallstudien untersucht der Beitrag exemplarische Konfigurationen einer investigativen Architekturproduktion, um aufzuzeigen, welche raumkonstituierenden Verfahren der Evidenzerzeugung entwickelt und eingesetzt werden, welche Authentifizierungsstrategien sie prägen und worin sich ein hegemoniekritischer von einem staatlich-repressiven Bildgebrauch unterscheidet. Damit verbunden ist die Frage nach dem Status der virtuellen Rekonstruktion im Spannungsfeld von Zeugnis und Beweis, nach ihrem Wert als Erkenntnisquelle und ihrer politischen wie ethischen Dimension. Nicht zuletzt geht es um die paradox anmutende Frage, wie digitale Raumbilder trotz ihrer Konstruiertheit und Manipulierbarkeit eine realitätsbeglaubigende Kraft in der investigativen Arbeit entfalten können.
