Zum Quellenwert deutscher Sprachzeugnisse aus Südosteuropa für die Sprachgeschichte des Deutschen

The history of the German language is not described adequately by the classification into Old, Middle High, Early New and New High German. Looking at regional varieties shows the simultaneous existence of older and newer manifestations in the language. Examples of older Transsylvanian writings and Danube-Swabian dialects from Romania and Hungary show how German varieties of South-Eastern Europe can be used for a more profound understanding of the history of the entire German language.
Die Publikation ornithologischer Daten setzt heute voraus,
dass sie mit angemessenen statistischen Methoden
ausgewertet werden. Mit der Entwicklung entsprechender
Verfahren steigen auch die Ansprüche an die
Auswertungen und an die Software, die für solche Auswertungen
nutzbar ist. Anerkannte kommerzielle Statistiksoftware
ist für den Normalverbraucher oft unerschwinglich
teuer. Das freie Statistikpaket R bietet eine
kostenlose, aber doch professionelle Lösung. Leider ist
der Einstieg in R nicht einfach, da das Programm nicht
geklickt werden kann, sondern Code geschrieben werden
muss. In diesem Artikel bieten wir eine Einstiegshilfe.
Wir zeigen Schritt für Schritt, wie mit R gearbeitet
wird. Der Leser kann direkt am eigenen Computer
nachvollziehen, wie Daten in das R eingelesen werden,
wie diese angesprochen und dargestellt werden. Wir
begleiten den Leser durch einen t-Test und führen ein
einfaches lineares Modell inklusive Residuenanalyse
durch. Abschließend geben wir Empfehlungen für weiterführende
Bücher.
A new, two-channel instrument for simultaneous NO3 and N2O5 monitoring was used to make the first comprehensive set of nocturnal NOx measurements (NO, NO2, NO3 and N2O5) at the Taunus Observatory, a rural mountain site (Kleiner Feldberg) in South-western Germany. In May 2008, NO3 and N2O5 mixing ratios were well above the instrumental detection limit (a few ppt) on all nights of the campaign and were characterised by large variability resulting from inhomogeneously distributed sinks. The concentrations of NO3, N2O5 and NO2 were consistent with the equilibrium constant, K2, defining the rates of formation and thermal dissociation of N2O5. A steady-state lifetime analysis showed that nocturnal NOx losses were generally dominated by reaction of NO3 with volatile organic compounds in this forested region, with N2O5 uptake to aerosols of secondary importance. Analysis of a limited dataset obtained at high relative humidity indicated that the loss of N2O5 by reaction with water vapour is less efficient (> factor 3) than derived using laboratory kinetic data. The fraction of NOx present as NO3 and N2O5 reached ≈20% on some nights, with night-time losses of NOx competing with daytime losses.
Am 20. Oktober 1781 wird das neue Theater in der Leopoldstadt (das "Kasperltheater") eröffnet, und es dauert nicht lange, bis Kasperl zur tragenden Figur der Bühne wird, die auch schnell den Spielplan prägt. La Roche betritt in jedem Monat etwa fünfzehnmal als Kasperl die Bühne und beherrscht diese fortan für viele Jahre. Ungeachtet der anderen Komiker, die ihr Glück als Kasperl versuchen, die aber tatsächlich nicht in der Lage waren, La Roches Ausdrucksweise zu erreichen, gelang es La Roche, sich förmlich in die erste Reihe der Wiener Vorstadtkomiker "zu spielen". Ein Weiterleben der Figur wurde nach La Roches Tod ebenso wie eine Renaissance unmöglich; auch auf anderen Theatern mussten die Akteure, die sich der "längst stereotyp gewordenen Mätzchen" befleißigten, und die Theaterdichter einsehen, dass eine das Repertoire derart bestimmende Typenkomik mit La Roche zu Grabe getragen worden war. Die Wirkung, welche die Person und vor allem das Spiel Johann Josef La Roches auf sein Publikum ausübte, wird in vielerlei Quellen beschworen: Zeitgenössische Kritiken stehen neben Erinnerungen von Schriftstellern, Chronisten sowie Biografen und geben ein vielfach beschworenes Bild, wie es La Roche gelang, seine Zuschauer zu fesseln. So illustrativ diese Erinnerungen auch sein mögen, so vermitteln sie doch ein rein subjektives Bild der Darstellungskunst La Roches. Pamphlete und Erwiderungen gibt es zuhauf, der Kasperl spaltete die Gemüter, Lob auf der einen, Verständnislosigkeit und vehemente Kritik auf der anderen Seite prägen mitunter die Rezensionen in den Zeitungen und auch einige der Broschüren Wiens. Ein wesentlicher Teil der Komik La Roches rührt von den Extempores her, steht Kasperl ja in der hanswurstischen Tradition. Auch wenn die "Anreden eines Schauspielers an das Parterre […] auch auf kleinen Volkstheatern […] nicht immer wohl schicklich" sind, so sollen La Roches Anspielungen auf Stadtereignissen gerade in seinen frühen Jahren "einen besonderen Reiz seines Spieles ausgemacht haben, obwohl es natürlich Gegner gab, die sich über 'Grobheiten und antastende Worte' entrüsteten". Die Wienerische Kronik spricht von "launichte[n] Einfälle[n]", die einer "gewissen schalkhaften Feinheit" nicht entbehren. Karl Marinelli engagierte Dichter, die eigens für den Wiener Kasperl Johann Josef La Roche Komödien verfassten. Schon sein Aussehen bewog das Publikum zu lachen, seine Sprache tat ein Übriges und vor allem die Verbindung von Sprache mit Spiel, Gestik und Mimik, Unausgesprochenem und Dargestelltem schien den besonderen Reiz auszumachen, mit dem La Roche das Publikum jahrzehntelang begeisterte. Leider sind die Berichte über La Roches darstellerischen Esprit auf Anekdoten und kaum verifizierbare Aussagen in Broschüren oder Chroniken beschränkt – für eine weitere Analyse muss auf die erhaltenen Textgrundlagen zurückgegriffen werden: auf Szenare und Kanevasse, auf die textlich fixierten Spielgrundlagen, deren Ausgestaltung durch die Sänger und Schauspieler nur erahnt werden kann. Aus den 30 im Projekt "Mäzene des Kasperls Johann Josef La Roche. Kasperliaden im Repertoire des Leopoldstädter Theaters. Kritische Edition und literatursoziologische Verortung" (2008/09) edierten Kasperliaden wurden vier hinsichtlich ihrer Kasperl-Komik analysiert. Die Auswahl erfolgte einerseits durch die Präsenz der Kasperlrolle im jeweiligen Text sowie andererseits durch die ihr je intendierte Relevanz für die komische Handlung (Qualität und Quantität der Kasperlrolle). Kasperl, der als Kaspar, Kasperl oder auch Käsperle auf die Bühne tritt, ist stets und in allen Stücken der Leopoldstädter Bühne als "besondere Person" gekennzeichnet – nicht durch seine ständische Zugehörigkeit oder durch seinen Beruf, sondern durch sein Verhalten, das ihn zum ersten von den anderen Rollen klar unterscheidet und das er gleichzeitig und zum zweiten dem Publikum gegenüber zeigt. Immer wieder fällt Kasperl aus der Rolle und durchbricht die Bühnenillusion für lustige Zwischenbemerkungen, um die Handlung zu kommentieren oder gar zu hinterfragen und um die Protagonisten zu bewerten. Die bemerkenswerteste "Entwicklung" durchläuft Kasperl im Oeuvre Karl Friedrich Henslers. Dabei avanciert die Figur vom rührigen und meist redlichen Familienvater in den bürgerlichen Stücken zum verschlagenen und bramarbasierenden Knappen in den romantisch-komischen Volksmärchen, als deren "Vater" Hensler in die Literatur- und Theatergeschichtsschreibung eingegangen ist. Dabei verläuft die Metamorphose vom bürgerlichen Biedermann zum komischen Faktotum in märchenhafter Kulisse in die eigentliche Handlung stets begleitenden Rollen, obschon Kasperl in vielen Fällen die Titel gebende Person ist. Der Kasperltypus wandelt sich merklich, doch immer bleibt er dumm, feige aber auch grundehrlich - damit sind seine Haupteigenschaften umrissen, die ihn in allen Komödien "begleiten". Der Furchtsamkeit, wie auch der Infantilität, die sich die Kasperlrolle im Laufe der Entwicklung beibehalten hatte, gesellt sich in den Volksmärchen ein Lazzo hinzu, nämlich der des Nachäffens, der typisch für den Kasperl La Roches wird.
Zwischen 1790, als Josef II. verstarb und Hoffnungen auf eine Lockerung der Zensur bestanden, und La Roches Todesjahr 1806 präsentierte das Leopoldstädter Theater in schneller Abfolge eine Unzahl von Possen, Singspielen, Maschinenkomödien, Zauberopern, Volksmärchen, Pantomimen, Ritterstücken, Soldatenstücken, Tanzspielen, Feenmärchen und komischen Zeitstücken. Sie stammten zu einem Gutteil von den Hausautoren des Theaters. [...] Aus den insgesamt 30 im Rahmen des FWF-Projekts "Mäzene des Kasperls Johann Josef La Roche" edierten Komödien von fünf Autoren und einem Anonymus wählt Beatrix Müller-Kampel zwei von jedem Autor aus, um sie auf Themen und Motive der Affekte, der Emotionen und ihrer Kontrolle hin zu durchforsten. Ausnahmslos alle Stücke sind Komödien, ausnahmslos allen geht es um Liebesgeschichten und Heiratssachen und den meisten auch um Geld. Das Prinzip der repräsentativen Stichprobe rechtfertigt sich durch die Gattung Komödie, wie sie am Leopoldstädter Theater gepflegt wurde: nämlich als "Schema-Dramatik", die im Gegensatz zur sogenannten "künstlerischen" Dramatik, d. h. in den theatralen Feldern dieser Zeit: im Gegensatz zum Bildungstheater einerseits, zum Hoftheater andrerseits, weder auf ästhetische Innovation noch auf Originalität abgestellt war, sondern im Gegenteil Variationen altbekannter Themen und Geschichten bieten wollte - und das immer auch komisch drapiert. [...] Mit dem Aspekt der Affekte und der Affektkontrolle der Lustigen Figur ist das Erkenntnisperspektiv einerseits auf Liebesgeschichten und Heiratssachen, Sich Verlieben wie Verlassen Werden, auf gelebte und gespielte Leidenschaften eingestellt, andrerseits auf die Lustige Figur und ihre Komik, welche letztere, so steht zu vermuten, in den Spielen wohl die großen wie die kleinen Gefühle modelliert. [...] Prinzipiell sind zwei Möglichkeiten der philologischen Emotionsforschung zu unterscheiden: die Analyse von "literarischen Thematisierungen und Darstellungen von Emotionen, wobei es in der Regel um Emotionen geht, die in einem Text irgendwelchen Figuren oder personifizierten Gegenständen zugeschrieben werden", und zweitens, die "historische Rekonstruktion kultureller Bewertungen und Repräsentationsformen diverser Emotionen." Was die dramatisch-sprachliche Darstellung von Emotionen anlangt, stehen in der Emotionsforschung mediale und mentalitätsgeschichtliche Aspekte im Mittelpunkt. Worin bestehen sie nun, die Emotionen, die sich zwischen Kasperl und den Frauen um 1800 auf dem Leopoldstädter Theater entspinnen, sich steigern, ausbreiten und verflüchtigen, und mit welchen dramaturgischen Techniken führen die Theaterautoren diese Emotionen dem Publikum vor? Kasperl, so das überraschende Fazit vorweg, ist um 1800 Hagestolz oder treuer Ehemann geworden. [...] Jedenfalls erstaunt Kasperls narrative, dramaturgische und komödiantische Bedeutungslosigkeit im Leopoldstädter Repertoire um 1800 - das doch für ihn und um ihn herum geschrieben worden war, wie es heißt. Wie, wenn mit seinen Affekten und seinem Affekthaushalt, das heißt beim Kasperl: mit seiner Obszönität, sich die Komik verflüchtigt und die Autoren tatsächlich nicht mehr gewusst hätten, was anzufangen sei mit ihm? In dem Maße, wie der Kasperl kein Frauensammler und Sexualphantast mehr sein darf, kein Zotenreißer und Hosenscheißer wie Hanswurst, kommt ihm textlich auch das Komische abhanden, das wohl tatsächlich ganz prinzipiell von einem lebt: dem Bruch von und dem Spiel mit Tabus.
Background: Decoding of frequency-modulated (FM) sounds is essential for phoneme identification. This study investigates selectivity to FM direction in the human auditory system. Methodology/Principal Findings: Magnetoencephalography was recorded in 10 adults during a two-tone adaptation paradigm with a 200-ms interstimulus-interval. Stimuli were pairs of either same or different frequency modulation direction. To control that FM repetition effects cannot be accounted for by their on- and offset properties, we additionally assessed responses to pairs of unmodulated tones with either same or different frequency composition. For the FM sweeps, N1m event-related magnetic field components were found at 103 and 130 ms after onset of the first (S1) and second stimulus (S2), respectively. This was followed by a sustained component starting at about 200 ms after S2. The sustained response was significantly stronger for stimulation with the same compared to different FM direction. This effect was not observed for the non-modulated control stimuli. Conclusions/Significance: Low-level processing of FM sounds was characterized by repetition enhancement to stimulus pairs with same versus different FM directions. This effect was FM-specific; it did not occur for unmodulated tones. The present findings may reflect specific interactions between frequency separation and temporal distance in the processing of consecutive FM sweeps.
Background: Diseases associated with smoking are a foremost cause of premature death in the world, both in developed and developing countries. Eliminating smoking can do more to improve health and prolong life than any other measure in the field of preventive medicine. Today's medical students will play a prominent role in future efforts to prevent and control tobacco use.
Methods: A cross-sectional, self-administered, anonymous survey of fifth-year medical students in Berlin, Germany was conducted in November 2007. The study explored the prevalence of smoking among medical students. We assessed their current knowledge regarding tobacco dependence and the effectiveness of smoking cessation methods. Students' perceived competence to counsel smokers and promote smoking cessation treatments was also explored. Analyses were based on responses from 258 students (86.6% response rate).
Results: One quarter of the medical students surveyed were current smokers. The smoking rate was 22.1% among women, 32.4% among men. Students underestimated smoking-related mortality and the negative effect of smoking on longevity. A considerable number of subjects erroneously assumed that nicotine causes coronary artery disease. Students' overall knowledge of the effectiveness of smoking cessation methods was inadequate. Only one third of the students indicated that they felt qualified to counsel patients about tobacco dependence.
Conclusions: This study reveals serious deficiencies in knowledge and counseling skills among medical students in our sample. The curriculum of every medical school should include a tobacco module. Thus, by providing comprehensive training in nicotine dependence interventions to medical students, smokers will have access to the professional expertise they need to quit smoking.

Background: Occupational demands of educators are not very well researched. Nevertheless their work is subject to several requirements. Whether these demands have an effect on the work ability and the health status of employees has also not been examined. Furthermore it is unclear if the ownership type of day care centres have an influence on job satisfaction and work ability of the pedagogical staff and what kind of resources do exist. Previous studies were mainly based on questionnaire data. Objective data does not exist. Therefore the aim of this investigation is to collect precise data relating to work of educators.
Methods: Effects of different types of ownership of day care centres on job satisfaction and work ability of educators will be assessed with the help of objective real time studies in combination with multi-level psycho diagnostic measurements.
Discussion: The present study is the first of its kind. Up to now there are no computer-based real time studies on workflow of pedagogical staff with regard to assess their work-related stress. Following an exhaustive documentation of educators work processes the day-to-day task can be estimated and approaches for prevention can be developed. This can substantially contribute to an overall improvement of child care in Germany.
Activation of Notch1 signaling in neural progenitor cells (NPCs) induces self-renewal and inhibits neurogenesis. Upon neuronal differentiation, NPCs overcome this inhibition, express proneural genes to induce Notch ligands, and activate Notch1 in neighboring NPCs. The molecular mechanism that coordinates Notch1 inactivation with initiation of neurogenesis remains elusive. Here, we provide evidence that Prox1, a transcription repressor and downstream target of proneural genes, counteracts Notch1 signaling via direct suppression of Notch1 gene expression. By expression studies in the developing spinal cord of chick and mouse embryo, we showed that Prox1 is limited to neuronal precursors residing between the Notch1+ NPCs and post-mitotic neurons. Physiological levels of Prox1 in this tissue are sufficient to allow binding at Notch1 promoter and they are critical for proper Notch1 transcriptional regulation in vivo. Gain-of-function studies in the chick neural tube and mouse NPCs suggest that Prox1-mediated suppression of Notch1 relieves its inhibition on neurogenesis and allows NPCs to exit the cell cycle and differentiate. Moreover, loss-of-function in the chick neural tube shows that Prox1 is necessary for suppression of Notch1 outside the ventricular zone, inhibition of active Notch signaling, down-regulation of NPC markers, and completion of neuronal differentiation program. Together these data suggest that Prox1 inhibits Notch1 gene expression to control the balance between NPC self-renewal and neuronal differentiation.
Objective: To summarise the benefits and harms of treatments for women with gestational diabetes mellitus. Design: Systematic review and meta-analysis of randomised controlled trials. Data sources: Embase, Medline, AMED, BIOSIS, CCMed, CDMS, CDSR, CENTRAL, CINAHL, DARE, HTA, NHS EED, Heclinet, SciSearch, several publishers’ databases, and reference lists of relevant secondary literature up to October 2009. Review methods: Included studies were randomised controlled trials of specific treatment for gestational diabetes compared with usual care or "intensified" compared with "less intensified" specific treatment. Results: Five randomised controlled trials matched the inclusion criteria for specific versus usual treatment. All studies used a two step approach with a 50 g glucose challenge test or screening for risk factors, or both, and a subsequent 75 g or 100 g oral glucose tolerance test. Meta-analyses did not show significant differences for most single end points judged to be of direct clinical importance. In women specifically treated for gestational diabetes, shoulder dystocia was significantly less common (odds ratio 0.40, 95% confidence interval 0.21 to 0.75), and one randomised controlled trial reported a significant reduction of pre-eclampsia (2.5 v 5.5%, P=0.02). For the surrogate end point of large for gestational age infants, the odds ratio was 0.48 (0.38 to 0.62). In the 13 randomised controlled trials of different intensities of specific treatments, meta-analysis showed a significant reduction of shoulder dystocia in women with more intensive treatment (0.31, 0.14 to 0.70). Conclusions: Treatment for gestational diabetes, consisting of treatment to lower blood glucose concentration alone or with special obstetric care, seems to lower the risk for some perinatal complications. Decisions regarding treatment should take into account that the evidence of benefit is derived from trials for which women were selected with a two step strategy (glucose challenge test/screening for risk factors and oral glucose tolerance test).
Background: European robins, Erithacus rubecula, show two types of directional responses to the magnetic field: (1) compass orientation that is based on radical pair processes and lateralized in favor of the right eye and (2) so-called 'fixed direction' responses that originate in the magnetite-based receptors in the upper beak. Both responses are light-dependent. Lateralization of the 'fixed direction' responses would suggest an interaction between the two magnetoreception systems. Results: Robins were tested with either the right or the left eye covered or with both eyes uncovered for their orientation under different light conditions. With 502 nm turquoise light, the birds showed normal compass orientation, whereas they displayed an easterly 'fixed direction' response under a combination of 502 nm turquoise with 590 nm yellow light. Monocularly right-eyed birds with their left eye covered were oriented just as they were binocularly as controls: under turquoise in their northerly migratory direction, under turquoise-and-yellow towards east. The response of monocularly left-eyed birds differed: under turquoise light, they were disoriented, reflecting a lateralization of the magnetic compass system in favor of the right eye, whereas they continued to head eastward under turquoise-and-yellow light. Conclusion: 'Fixed direction' responses are not lateralized. Hence the interactions between the magnetite-receptors in the beak and the visual system do not seem to involve the magnetoreception system based on radical pair processes, but rather other, non-lateralized components of the visual system.
Vascularization of the vertebrate brain takes place during embryonic development from a preformed perineural vascular plexus. As a consequence of the intimate contact with neuroectodermal cells the vessels, which are entering the brain exclusively via sprouting angiogenesis, acquire and maintain unique barrier properties known as the blood-brain barrier (BBB). The endothelial BBB depends upon the close association of endothelial cells with pericytes, astrocytes, neurons and microglia, which are summarized in the term neuro-vascular unit. Although it is known since decades that the CNS tissue provides the cues for BBB induction and differentiation in endothelial cells, the molecular mechanism remained obscure. Only recently, the canonical Wnt/beta-catenin pathway and the Wnt7a/7b growth factors have been implicated in brain angiogenesis on the one hand and in BBB induction on the other. This breakthrough in understanding the differentiation of the brain vasculature prompted us to review these findings embedded in the emerging concepts of Wnt signaling in the vasculature. In particular, interactions with other pathways that are crucial for vascular development such as VEGF, Notch, angiopoietins and Sonic hedgehog are discussed. Finally, we considered the potential role of the Wnt pathway in vascular brain pathologies in which BBB function is hampered, as for example in glioma, stroke and Alzheimer's disease.
A comprehensive evaluation of seasonal backward trajectories initialized in the Northern Hemisphere lowermost stratosphere (LMS) has been performed to investigate the origin of air parcels and the main mechanisms determining characteristic structures in H2O and CO within the LMS. In particular we explain the fundamental role of the transit time since last tropopause crossing (tTST) for the chemical structure of the LMS as well as the feature of the extra-tropical tropopause transition layer (ExTL) as identified from CO profiles. The distribution of H2O in the background LMS above Θ=320 K and 340 K in northern winter and summer, respectively, is found to be governed mainly by the saturation mixing ratio, which in turn is determined by the Lagrangian Cold Point (LCP) encountered by each trajectory. Most of the backward trajectories from this region in the LMS experienced their LCP in the tropics and sub-tropics. The transit time since crossing the tropopause from the troposphere to the stratosphere (tTST) is independent of the H2O value of the air parcel. TST often occurs 20 days after trajectories have encountered their LCP. CO, on the other hand, depends strongly on tTST due to its finite lifetime. The ExTL as identified from CO measurements is then explained as a layer of air just above the tropopause, which on average encountered TST fairly recently.
Blood vessels form de novo through the tightly regulated programs of vasculogenesis and angiogenesis. Both processes are distinct but one of the steps they share is the formation of a central lumen, when groups of cells organized as vascular cords undergo complex changes to achieve a tube-like morphology. Recently, a protein termed epidermal growth factor-like domain 7 (EGFL7) was described as a novel endothelial cell-derived factor involved in the regulation of the spatial arrangement of cells during vascular tube assembly. With its impact on tubulogenesis and vessel shape EGFL7 joined the large family of molecules governing blood vessel formation. Only recently, the molecular mechanisms underlying EGFL7's effects have been started to be elucidated and shaping of the extracellular matrix (ECM) as well as Notch signaling might very well play a role in mediating its biological effects. Further, findings in knock-out animal models suggest miR-126, a miRNA located within the egfl7 gene, has a major role in vessel development by promoting VEGF signaling, angiogenesis and vascular integrity. This review summarizes our current knowledge on EGFL7 and miR-126 and we will discuss the implications of both bioactive molecules for the formation of blood vessels.
A comprehensive evaluation of seasonal backward trajectories initialized in the northern hemisphere lowermost stratosphere (LMS) has been performed to investigate the factors that determine the temporal and spatial structure of troposphere-to-stratosphere-transport (TST) and it’s impact on the LMS. In particular we explain the fundamental role of the transit time since last TST (tTST) for the chemical composition of the LMS. According to our results the structure of the LMS can be characterized by a layer with tTST<40 days forming a narrow band around the local tropopause. This layer extends about 30K above the local dynamical tropopause, corresponding to the extratropical tropopause transition layer (ExTL) as identified by CO. The LMS beyond this layer shows a relatively well defined separation as marked by an aprupt transition to longer tTST indicating less frequent mixing and a smaller fraction of tropospheric air. Thus the LMS constitutes a region of two well defined regimes of tropospheric influence. These can be characterized mainly by different transport times from the troposphere and different fractions of tropospheric air. Carbon monoxide (CO) mirrors this structure of tTST due to it’s finite lifetime on the order of three months. Water vapour isopleths, on the other hand, do not uniquely indicate TST and are independent of tTST, but are determined by the Lagrangian Cold Point (LCP) of air parcels. Most of the backward trajectories from the LMS experienced their LCP in the tropics and sub-tropics, and TST often occurs 20 days after trajectories have encountered their LCP. Therefore, ExTL properties deduced from CO and H2O provide totally different informations on transport and particular TST for the LMS.

This paper discusses the sustainability impact (contribution to sustainability, reduction of adverse environmental impacts) of online second-hand trading. A survey of eBay users shows that a relationship between the trading of used goods and the protection of natural resources is hardly realized. Secondly, the environmental motivation and the willingness to act in a sustainable manner differ widely between groups of consumers. Given these results from a user perspective, the paper tries to find some objective hints of online second-hand trading’s environmental impact. The greenhouse gas emissions resulting from the energy used for the trading transactions seem to be considerably lower than the emissions due to the (avoided) production of new goods. The paper concludes with a set of recommendations for second-hand trade and consumer policy. Information about the sustainability benefits of purchasing second-hand goods should be included in general consumer information, and arguments for changes in behavior should be targeted to different groups of consumers. In den Vorlesungen zur Gouvernementalität skizziert Foucault die Art und Weise, in der im modernen Staat «aus der Distanz» regiert wird. Diese wird im Artikel dargestellt, materialistisch «geerdet», und es werden hierauf aufbauend die Begriffe Risiko und Versicherheitlichung diskutiert. Die Tauglichkeit dieser Herangehensweise wird anhand der aktuellen Grenz- und Migrationspolitik der EU illustriert, und es werden die in diesem Kontext produzierten Räume skizziert.
Erzählt wurde und wird überall. Erzählen ist eine Grundform unserer Wirklichkeitserfassung. Das Zentrum für Erzählforschung (ZEF) der Bergischen Universität ist deshalb interdisziplinär ausgerichtet. Drei Beispiele aus Kunstgeschichte, Journalismus und Rechtsprechung: Der Teppich von Bayeux (11. Jh.) benutzt bei seiner bildlichen Darstellung der Eroberung Englands durch den Normannen Wilhelm Abweichungen von der Chronologie, um die Sympathien der Betrachter zu lenken. Die pietätlose Hast des angelsächsischen Kontrahenten der Normannen, Harold, sich nach dem Tod seines Vorgängers krönen zu lassen, wird visuell durch die unmittelbare Konfrontation der Krönungsszene mit dem Bild des kranken Edward angezeigt. In solchen erzählerischen Kunstgriffen zeigt sich politische Wirkungsabsicht. Zweitens: Fußballspiele stellen den Live-Reporter vor große erzählerische Herausforderung. Er muss eine Geschichte erzählen, deren Ende er nicht kennt. Dafür verwendet er prospektiv Plotstrukturen, die dem disparaten Spielgeschehen Sinn verleihen, die aber auch gegebenenfalls ad hoc abweichendem Spielverlauf anzupassen sind. Drittens: Vor Gericht wird die Glaubhaftigkeit von Zeugenaussagen u. a. nach erzählerischen Kriterien wie Detaillierungsgrad und Inkontinenz beurteilt. Wer besser erzählt, gewinnt.
1987 überraschte der chinesische Künstler Xu Bing sein Pekinger Publikum mit einem Set von vier Büchern, die im Stil der kanonischen Bücher gestaltet waren und über tausendzweihundert chinesische Schriftzeichen enthielten. Obwohl auf den ersten Blick den alten chinesischen Zeichen täuschend ähnlich, unterschieden sie sich jedoch von ihnen in einigen wenigen Details, die entscheidend für das Verständnis waren. Xu Bings Idee bestand darin, einem alten Wörterbuch reale Zeichen zu entnehmen, sie in ihre Grundbestandteile zu zerlegen und sie wieder zusammensetzen, jedoch nicht so, wie sie vorher waren, sodass sie ihrer Bedeutung beraubt wurden. Angesichts dieser gekonnt inszenierten und verwirrenden Lektüreerfahrung stellt sich die Frage: Ist das überhaupt Literatur, wo es doch nichts zu lesen gibt? Diese Frage mag ungewöhnlich klingen und nicht wenige werden sie vielleicht sogar für falsch gestellt halten. Schließlich gilt Xu Bing als Künstler und nicht als Schriftsteller und seine ausgestellten Bücher sind in dem Ausstellungsraum eines Museums zu finden und nicht in einer Bibliothek. Und dennoch, so die Hypothese dieser Überlegungen, findet man in seinen Büchern etwas zur Darstellung gebracht, was nach Foucault so etwas ist wie der unsagbare Kern der Literatur. In Xu Bings Ausstellung geht es um die sichtbare Manifestation dessen, was Literatur im sprachontologischen Sinne begründet, um ihre Existenzbedingung: das Nichtsignifikative im Zeichen, das die Signifikation erst ermöglicht. In Analogie zu Blanchots Begriff "desoeuvrement" (vgl. Blanchot 1955, 48 f) könnte man von einer 'désignification' sprechen: Durch winzige Veränderungen am Gefüge der Striche wird dem Zeichen der Sinn entzogen. Doch wie lässt sich der Bezug dieses unlesbaren Buches, das aufgrund seiner Unlesbarkeit nur noch als Ausstellungsobjekt taugt, zum literarischen Buch herstellen?
Tracer measurements in the tropical tropopause layer during the AMMA/SCOUT-O3 aircraft campaign

We present airborne in situ measurements made during the AMMA (African Monsoon Multidisciplinary Analysis)/SCOUT-O3 campaign between 31 July and 17 August 2006 on board the M55 Geophysica aircraft, based in Ouagadougou, Burkina Faso. CO<sub>2</sub> and N<sub>2</sub>O were measured with the High Altitude Gas Analyzer (HAGAR), CO was measured with the Cryogenically Operated Laser Diode (COLD) instrument, and O<sub>3</sub> with the Fast Ozone ANalyzer (FOZAN). We analyze the data obtained during five local flights to study the dominant transport processes controlling the tropical tropopause layer (TTL) above West-Africa: deep convection up to the level of main convective outflow, overshooting of deep convection, horizontal inmixing across the subtropical tropopause, and horizontal transport across the subtropical barrier. Except for the flight of 13 August, distinct minima in CO<sub>2</sub> indicate convective outflow of boundary layer air in the TTL. The CO<sub>2</sub> profiles show that the level of main convective outflow was mostly located between 350 and 360 K, and for 11 August reached up to 370 K. While the CO<sub>2</sub> minima indicate quite significant convective influence, the O<sub>3</sub> profiles suggest that the observed convective signatures were mostly not fresh, but of older origin. When compared with the mean O<sub>3</sub> profile measured during a previous campaign over Darwin in November 2005, the O<sub>3</sub> minimum at the main convective outflow level was less pronounced over Ouagadougou. Furthermore O<sub>3</sub> mixing ratios were much higher throughout the whole TTL and, unlike over Darwin, rarely showed low values observed in the regional boundary layer. Signatures of irreversible mixing following overshooting of convective air were scarce in the tracer data. Some small signatures indicative of this process were found in CO<sub>2</sub> profiles between 390 and 410 K during the flights of 4 and 8 August, and in CO data at 410 K on 7 August. However, the absence of expected corresponding signatures in other tracer data makes this evidence inconclusive, and overall there is little indication from the observations that overshooting convection has a profound impact on TTL composition during AMMA. We find the amount of photochemically aged air isentropically mixed into the TTL across the subtropical tropopause to be not significant. Using the N<sub>2</sub>O observations we estimate the fraction of aged extratropical stratospheric air in the TTL to be 0.0±0.1 up to 370 K during the local flights, increasing above this level to 0.2±0.15 at 390 K. The subtropical barrier, as indicated by the slope of the correlation between N<sub>2</sub>O and O<sub>3</sub> between 415 and 490 K, does not appear as a sharp border between the tropics and extratropics, but rather as a gradual transition region between 10 and 25° N latitude where isentropic mixing between these two regions may occur.

'One Plus One' entstand während einer über mehrere Monate verteilten Aufnahmestudio-Session der Rolling Stones im Juni und August 1968 in den Olympic Studios in London. Die Band kam ohne Vorbereitung ins Studio und entwickelte den Song 'Sympathy for the Devil' improvisatorisch im Laufe der Sessions. Die Arbeit am Stück begann mit einem Liedtext von Sänger Mick Jagger unter dem Arbeitstitel 'The Devil Is My Name'. Jagger hatte diesen Text unter dem Eindruck des Romans 'Der Meister und Margarita' des russischen Schriftstellers Michail Bulgakov verfasst - ein Buch, das er von seiner Partnerin Marianne Faithfull erhalten hatte. In Bulgakovs Roman stattet der Teufel dem Moskau der 1930er Jahre einen Besuch ab, in dessen Verlauf er viele Menschen tötet oder in den Wahnsinn treibt. Der Film zeigt die Arbeit an dem Song, der auf einem treibenden Samba-Rhythmus aufruht, in den verschiedenen Entwicklungsstufen, in immer neuen Ansätzen, sich dem späteren Format annähernd. Der Song wurde 1968 auf dem Studioalbum 'Beggars Banquet' als erstes von zehn Liedern veröffentlicht.
Small interfering RNAs (siRNAs) are now established as the preferred tool to inhibit gene function in mammalian cells yet trigger unintended gene silencing due to their inherent miRNA-like behavior. Such off-target effects are primarily mediated by the sequence-specific interaction between the siRNA seed regions (position 2–8 of either siRNA strand counting from the 5'-end) and complementary sequences in the 3'UTR of (off-) targets. It was previously shown that chemical modification of siRNAs can reduce off-targeting but only very few modifications have been tested leaving more to be identified. Here we developed a luciferase reporter-based assay suitable to monitor siRNA off-targeting in a high throughput manner using stable cell lines. We investigated the impact of chemically modifying single nucleotide positions within the siRNA seed on siRNA function and off-targeting using 10 different types of chemical modifications, three different target sequences and three siRNA concentrations. We found several differently modified siRNAs to exercise reduced off-targeting yet incorporation of the strongly destabilizing unlocked nucleic acid (UNA) modification into position 7 of the siRNA most potently reduced off-targeting for all tested sequences. Notably, such position-specific destabilization of siRNA–target interactions did not significantly reduce siRNA potency and is therefore well suited for future siRNA designs especially for applications in vivo where siRNA concentrations, expectedly, will be low.
Hrad Wartburg v Durynsku je jedním z nejvýznamnějších památníků německé kultury. Jeho jméno je spojeno s dějinami minnesangu stejně jako s historií německé reformace. Ve Wagnerově opeře 'Tannhäuser' se však tento hrad stává především symbolem, metaforou, za níţ se skrývá tradiční německá "Kultura" (tak jak ji definoval např. Thomas Mann ve svých 'Betrachtungen eines Unpolitischen'). Novější německé dějiny znají hned několik epoch, v nichţ hrály hodnoty reprezentované wagnerovským Wartburgem významnou roli – wilhelmovská "Druhá" stejně jako – ještě radikálněji – Hitlerova "Třetí říše". Cílem tohoto příspěvku je objasnit na příkladu německé literatury, jak "wartburské" tyto doby byly a jak reálné je zrcadlo, které Wagner se svým 'Tannhäuserem' nastavil (německému) světu.
In vielen Familien der Mittelschicht sind mittlerweile beide Ehepartner berufstätig. Das männliche Alleinverdiener-Modell verschwindet zwar nicht, aber nimmt ab. Die Arbeit zu Hause ist trotzdem noch ungleich verteilt und überwiegend Frauensache geblieben – wenn auch häufig unter neuen Vorzeichen: Oft übernehmen Migrantinnen, insbesondere aus Osteuropa, einen Teil der Care-Arbeit. Welche Konsequenzen hat dies für deren Familien?
We synthesised observations of total particle number (CN) concentration from 36 sites around the world. We found that annual mean CN concentrations are typically 300–2000 cm -3 in the marine boundary layer and free troposphere (FT) and 1000–10 000 cm -3 in the continental boundary layer (BL). Many sites exhibit pronounced seasonality with summer time concentrations a factor of 2–10 greater than wintertime concentrations. We used these CN observations to evaluate primary and secondary sources of particle number in a global aerosol microphysics model. We found that emissions of primary particles can reasonably reproduce the spatial pattern of observed CN concentration (R2=0.46) but fail to explain the observed seasonal cycle (R2=0.1). The modeled CN concentration in the FT was biased low (normalised mean bias, NMB=& -88%) unless a secondary source of particles was included, for example from binary homogeneous nucleation of sulfuric acid and water (NMB= -25%). Simulated CN concentrations in the continental BL were also biased low (NMB= -74%) unless the number emission of anthropogenic primary particles was increased or a mechanism that results in particle formation in the BL was included. We ran a number of simulations where we included an empirical BL nucleation mechanism either using the activation-type mechanism (nucleation rate, J, proportional to gas-phase sulfuric acid concentration to the power one) or kinetic-type mechanism (J proportional to sulfuric acid to the power two) with a range of nucleation coefficients. We found that the seasonal CN cycle observed at continental BL sites was better simulated by BL particle formation (R2=0.3) than by increasing the number emission from primary anthropogenic sources (R2=0.18). The nucleation constants that resulted in best overall match between model and observed CN concentrations were consistent with values derived in previous studies from detailed case studies at individual sites. In our model, kinetic and activation-type nucleation parameterizations gave similar agreement with observed monthly mean CN concentrations.
Introduction: Researchers experience increasing pressures to connect with bodies that finance their projects. In this climate, critical scholars face many obstacles as they seek to navigate the treacherous waters of securing external funds. To debate these challenges, the ACME Editorial Collective organized a panel for the 2009 Annual Meeting of the Association of American Geographers in Las Vegas. This intervention represents a follow-up discussion and collective writing process among some of the panelists and members of the audience who attended the panel.
Below, we examine the neoliberalization of the current funding systems, discuss the implications for research practice, and make suggestions for critical engagement and transformation. Our suggestions, however, will not be easy to implement, as we can infer from the experience of the radical scholars of the post-1968 generation whose ascension into the upper echelons of North American and European university systems was also associated with the neoliberalization of the funding systems. This intervention represents a modest contribution in the tradition of critical research practice of creating the possibilities for progressive change.
Intracarotid administration of human bone marrow mononuclear cells in rat photothrombotic ischemia

Background: Increasing evidence suggests that cell therapy improves functional recovery in experimental models of stroke and myocardial infarction. So far only small pilot trials tested the effects of cell therapy in stroke patients, whereas large clinical trials were conducted in patients with ischemic heart disease. To investigate the therapeutic benefit of cell therapy to improve the recovery after stroke, we determined the efficacy of bone marrow derived mononuclear cells, which were shown to improve the recovery in experimental and clinical acute myocardial infarction studies, in a rat stroke model. Methods: Adult male Wistar rats were randomly assigned to receive either five million human bone marrow mononuclear cells (hBMC) or placebo intraarterially 3 days after photothrombotic ischemia. For immunosuppression the animals received daily injections of cyclosporine throughout the experiment, commencing 24 hours before the cell transplantation. A battery of behavioural tests was performed before and up to 4 weeks after ischemia. Results: Body temperature and body weight revealed no difference between groups. Neurological deficits measured by the Rotarod test, the adhesive-removal test and the cylinder test were not improved by hBMC transplantation compared to placebo. Conclusions: This study demonstrates that hBMC do not improve functional recovery when transplanted intraaterially 3 days after the onset of focal cerebral ischemia. A possible reason for the failed neurological improvement after cell therapy might be the delayed treatment initiation compared to other experimental stroke studies that showed efficacy of bone marrow mononuclear cells.

Romanian readers interested in Max Frisch’s literary diaries can find the Romanian translation of some heterogeneous notes or groups of notes, dispersed in different periodicals from 1972 to 2001, and the volume Journal (1984) including a selection of fragments from the diaries Tagebuch 1946-1949 and Tagebuch 1966-1971 translated by Corina Jiva. The present article emphasizes favorite as well as excluded thematic fields while making the selection and pinpoints the translator’s role as an authority endowed with preventive censorship.
Background: ClC-7 is a ubiquitous transporter which is broadly expressed in mammalian tissues. It is implied in the pathogenesis of lysosomal storage disease and osteopetrosis. Because of its endosomal/lysosomal localization it is still poorly characterized. Methodology/Principal Findings: An electrophysiological characterization of rat ClC-7 using solid-supported membrane-based electrophysiology is presented. The measured currents show the characteristics of ClC-7 and confirm its function as a Cl−/H+-antiporter. We have used rat ClC-7 in CHO cells as a model system to investigate the functionality and cellular localization of the wt transporter and its variant G213R ClC-7 which is the analogue of human G215R ClC-7 responsible for autosomal dominant osteopetrosis type II. Our study shows that rat G213R ClC-7 is functional but has a localization defect in CHO cells which prevents it from being correctly targeted to the lysosomal membrane. The electrophysiological assay is tested as a tool for drug discovery. The assay is validated with a number of drug candidates. It is shown that ClC-7 is inhibited by DIDS, NPPB and NS5818 at micromolar concentrations. Conclusions/Significance: It is suggested that the scenario found in the CHO model system also applies to the human transporter and that mislocalization rather than impaired functionality of G215R ClC-7 is the primary cause of the related autosomal dominant osteopetrosis type II. Furthermore, the robust solid-supported membrane-based electrophysiological assay is proposed for rapid screening for potential ClC-7 inhibitors which are discussed for treatment of osteoporosis.
The human immunodeficiency virus type 1 (HIV-1) coreceptor use and viral evolution were analyzed in blood samples from an HIV-1 infected patient undergoing allogeneic stem cell transplantation (SCT). Coreceptor use was predicted in silico from sequence data obtained from the third variable loop region of the viral envelope gene with two software tools. Viral diversity and evolution was evaluated on the same samples by Bayesian inference and maximum likelihood methods. In addition, phenotypic analysis was done by comparison of viral growth in peripheral blood mononuclear cells and in a CCR5 (R5)-deficient T-cell line which was controlled by a reporter assay confirming viral tropism. In silico coreceptor predictions did not match experimental determinations that showed a consistent R5 tropism. Anti-HIV directed antibodies could be detected before and after the SCT. These preexisting antibodies did not prevent viral rebound after the interruption of antiretroviral therapy during the SCT. Eventually, transplantation and readministration of anti-retroviral drugs lead to sustained increase in CD4 counts and decreased viral load to undetectable levels. Unexpectedly, viral diversity decreased after successful SCT. Our data evidence that only R5-tropic virus was found in the patient before and after transplantation. Therefore, blocking CCR5 receptor during stem cell transplantation might have had beneficial effects and this might apply to more patients undergoing allogeneic stem cell transplantation. Furthermore, we revealed a scenario of HIV-1 dynamic different from the commonly described ones. Analysis of viral evolution shows the decrease of viral diversity even during episodes with bursts in viral load.
Background: One virulence property of Borrelia burgdorferi is its resistance to innate immunity, in particular to complement-mediated killing. Serum-resistant B. burgdorferi express up to five distinct complement regulator-acquiring surface proteins (CRASP) which interact with complement regulator factor H (CFH) and factor H-like protein 1 (FHL1) or factor H-related protein 1 (CFHR1). In the present study we elucidate the role of the infection-associated CRASP-3 and CRASP-5 protein to serve as ligands for additional complement regulatory proteins as well as for complement resistance of B. burgdorferi. Methodology/Principal Findings: To elucidate whether CRASP-5 and CRASP-3 interact with various human proteins, both borrelial proteins were immobilized on magnetic beads. Following incubation with human serum, bound proteins were eluted and separated by Glycine-SDS-PAGE. In addition to CFH and CFHR1, complement regulators CFHR2 and CFHR5 were identified as novel ligands for both borrelial proteins by employing MALDI-TOF. To further assess the contributions of CRASP-3 and CRASP-5 to complement resistance, a serum-sensitive B. garinii strain G1 which lacks all CFH-binding proteins was used as a valuable model for functional analyses. Both CRASPs expressed on the B. garinii outer surface bound CFH as well as CFHR1 and CFHR2 in ELISA. In contrast, live B. garinii bound CFHR1, CFHR2, and CFHR5 and only miniscute amounts of CFH as demonstrated by serum adsorption assays and FACS analyses. Further functional analysis revealed that upon NHS incubation, CRASP-3 or CRASP-5 expressing borreliae were killed by complement. Conclusions/Significance: In the absence of CFH and the presence of CFHR1, CFHR2 and CFHR5, assembly and integration of the membrane attack complex was not efficiently inhibited indicating that CFH in co-operation with CFHR1, CFHR2 and CFHR5 supports complement evasion of B. burgdorferi.

Vereinzelt kommt es auch zu Übersommerungen.
Die Herkunft dieser Vögel ist weitgehend unbekannt.
Erstmals in Mitteleuropa konnte von uns ein
Silberreiher mit einem Satellitensender versehen und
von Februar 2007 bis zum Ende der Datenaufnahme
für diesen Bericht im April 2010 verfolgt werden. Der
Vogel überwinterte dreimal in Ostdeutschland und hielt
sich 2008 und 2009 während der Brutzeit sehr ortstreu

Wanderungen über mehrere 100 km, die als Mauserzug
interpretiert werden. Erst im Laufe des Oktobers
fand der eigentliche Zug ins Überwinterungsgebiet
statt. Ringfunde aus der Zeit nach dem deutlichen Bestandsanstieg
in Deutschland geben außerdem erste Hinweise darauf, dass die im Nordosten Deutschlands
als Gastvögel auftretenden Silberreiher auch aus Westfrankreich
und Polen stammen können und dass zumindest
in Süddeutschland Vögel aus den traditionellen
ungarischen Brutgebieten auftreten.
Die Ermittlung von Überlebenswahrscheinlichkeiten und Reproduktionsdaten hat eine wachsende Bedeutung als Basis für Populationsmodelle gewonnen. Wir haben Beringungen und Rückmeldungen aus dem Datenbestand der Beringungszentrale Hiddensee und die Ergebnisse aus einer Farbberingungsstudie an 110 Rotkehlchen Erithacus rubecula benutzt, um mit dem Programm MARK für vier häufige Zugvogelarten Überlebenswahrscheinlichkeiten zu schätzen und die Möglichkeiten und Grenzen bei der Nutzung der Datenbank einer Beringungszentrale aufzuzeigen. Es wurden je nach Struktur der Daten entweder Cormack-Jolly-Seber-Modelle (CJS-Modelle) für Wiederfänge und Ringablesungen oder Modelle nach Burnham (1993) für kombinierte Daten aus Totfunden und Wiederfängen verwendet. Für drei Arten konnten wir so plausible Überlebenswahrscheinlichkeiten ermitteln, es gab aber deutliche Unterschiede in der Präzision je nach Vogelart sowie verwendeter Datengrundlage. Bei Fang-Wiederfang-Daten beeinflusste nicht zu modellierende Abwanderung die Ergebnisse für mindestens eine Altersklasse, d. h. wenigstens ein Parameter konnte nicht geschätzt werden. Zudem waren eine ausreichend hohe Wiederfundwahrscheinlichkeit sowie eine vollständige Meldung der Wiederfänge wichtige Voraussetzungen für zuverlässige Ergebnisse. Sie wurden hier durch eine Vorauswahl von Datensätzen aus Gebieten mit mehrjähriger intensiver Fangaktivität erreicht. Kombinierte Modelle aus Totfunden und Wiederfängen waren leichter auf den vollständigen Datenbestand der Beringungszentrale und für alle Altersklassen anwendbar. Bei auf dem Zug oder im Winterquartier stark bejagten Arten, für die genügend Totfunde vorliegen, wurden so brauchbare Ergebnisse erzielt. Daten aus dem Fang von Durchzüglern enthielten teilweise zu wenig verwertbare Rückmeldungen, auch wenn jährlich große Mengen von Vögeln beringt werden („Registrierfang“).
Vegetation und Management seltener Pflanzenarten im Küstengrünland einer dänischen Ostseeinsel

Die vorliegende Arbeit beschreibt die Vegetation des Küstengrünlandes im Naturschutzgebiet „Vestamager“ (ca. 2000 ha) südlich von Kopenhagen sowie Vorkommen von Iris spuria, Selinum dubium und Cerastium subtetrandrum. Die mit diesen Arten verbundenen Pflanzengesellschaften und Standort - faktoren werden ermittelt, um das Management des Naturschutzgebietes zu verbessern. Auf der Grundlage von 73 Vegetationsaufnahmen werden mit TWINSPAN-Klassifikation und indirekter Gradientenanalyse (DCA) die Hauptvegetationseinheiten und wichtigsten Umweltgradienten bestimmt. Die Vegetationsentwicklung 2004–2008 wird in einer weiteren Ordination mit Hilfe historischer Aufnahmen untersucht. Die vegetationsbestimmenden Faktoren auf Vestamager sind Salinität, Feuchtigkeit und Beweidung. Magere, salzgeprägte Weidegesellschaften des Lolio-Cynosuretum sind häufig; innerhalb dieser Assoziation ist zwischen einer wechselfeuchten Ausbildung mit Molinion-Elementen und einer wechseltrockenen Ausbildung mit floristischen Beziehungen zum Mesobromium zu unterscheiden. Im Südwesten des Gebietes wird das Lolio-Cynosuretum mit steigender Salinität durch Salzrasen des Juncetum gerardii ersetzt. Innerhalb der Salzrasen ist auf Ameisenhaufen Salzpioniervegetation entwickelt. Der Übergangsbereich zum Hydrolitoral wird von Brackwasserröhrichten besiedelt. Auf feuchten bis nassen Standorten geht das Lolio-Cynosuretum in Flut- und Trittrasengesellschaften des Ranunculo repentis-Alopecuretum geniculati über. Bei schwacher Beweidung bilden sich Calamagrostis epigejos- Dominanzbestände aus. Iris spuria und Selinum dubium kommen mit je einer Population in der wechseltrockenen Ausprägung des Lolio-Cynosuretum juncetosum gerardii im nördlichen Teil des Gebietes vor. Kleinere Vorkommen von Selinum dubium finden sich auch in Flut- und Trittrasengesellschaften. Cerastium subtetrandrum kommt mit mehreren Kleinpopulationen in Salzpionierrasen auf Ameisenhaufen vor. Iris spuria reagiert empfindlich auf Beweidung und ist in den letzten Jahren deutlich zurückgegangen. Selinum dubium ist toleranter gegenüber Beweidung, und vegetative Individuen können auch auf beweideten Flächen überdauern. Cerastium subtetrandrum ist indirekt von Beweidung abhängig, da potentielle Standorte dieser Art durch Beweidung erhalten und gefördert werden. Intensivierte Beweidung sowie Entfernen von Gehölzen haben Hochgräser zurückgedrängt und Arten offener, salzgeprägter Weidelandschaften gefördert, jedoch ohne Erhöhung der floristischen Diversität. Der optimale Schutz der Zielarten kann nur durch kombinierte Nutzungskonzepte erreicht werden. Für eine langfristige Erhaltung von Iris spuria und Selinum dubium ist eine Reduzierung der Beweidung notwendig. Die Erhaltung von Cerastium subtetrandrum erfordert dagegen eine niedrige Vegetation mit relativ intensiver Beweidung. Eine stärkere Differenzierung der Beweidungsintensität würde alle drei Zielarten auf Vestamager fördern.

Zwei konstruierte Todesfälle gewähren einen forensischen Blick auf das Blut.

Die von Gabriele Mentges (Dortmund) und Gertrud Lehnert (Potsdam) veranstaltete und von der Volkswagen Stiftung geförderte Tagung befasste sich mit dem Thema 'Orientalismus und Mode' zum einen unter historischen Aspekten, zum anderen unter gegenwärtigen Bedingungen, in denen von 'Orientalismus' kaum noch die Rede sein kann. Denn längst haben wechselseitige Bezüge Mode zu einer globalen Angelegenheit gemacht. Vernetzung, Vermischung, Globalisierung, aber auch neue Regionalisierung bestimmt (nicht erst seit) heute die Mode.
In contrast to the previous structure determinations of the title structure, (NH4)2[MoS4], the present determination at 173 K localized the positions of the H atoms. The title structure belongs to the beta-K2SO4 family and all the ions are located on crystallographic mirror planes. The ions are held together by N—H ... S hydrogen bonds (some of which are bifurcated), forming a three-dimensional network. One of the N atoms has nine contacts to the S atoms shorter than 4 Å, and the other has ten.
The asymmetric unit of the title compound, [K(C5HF6N2)(H2O)2]n, is composed of two 3,5-bis(trifluoromethyl)pyrazolide anions, two potassium cations and four water molecules. The water molecules and 3,5-bis(trifluoromethyl)pyrazolide anions act as bridges between the potassium cations. Each potassium cation is surrounded by four O atoms [K—O = 2.705 (3)–2.767 (3) Å] and four F atoms [K—F = 2.870 (7)–3.215 (13) Å]. The water molecules and the 3,5-bis(trifluoromethyl)pyrazolide anions are connected by O—H ... N hydrogen bonds, forming layers in the ab plane. All –CF3 groups show rotational disorder between two orientations each.
The title compound, [Na(CF3O3S)(C12H24O6)], features a sodium cation that is coordinated by eight O atoms in an irregular hexagonal bipyramidal environment. The equatorial positions are occupied by the six O atoms of an 18-crown-6 ether ring. In the axial positions, there is one O atom of a trifluoromethanesulfonate anion and an ether O atom of a symmetry-equivalent crown ether ring. In this way, centrosymmetric dimers are formed.
The central structural element of the title compound, C24H29NO2, is a carbazole unit substituted with two acetyl residues and an octyl chain. The acetyl residues are nearly coplanar [dihedral angles = 5.37 (14) and 1.0 (3)°] with the carbazole unit which is essentially planar (r.m.s. deviation for all non-H atoms = 0.025 Å). The octyl chain adopts an all-trans conformation. The crystal packing is stabilized by C—H ... O hydrogen bonds.
The dihydropyrimidine ring of the title compound, C13H15ClN2S, adopts an envelope conformation with five almost coplanar atoms (r.m.s. deviation = 0.054 Å) and the C atom bearing the two methyl substituents deviating from this plane by 0.441 (2) Å. The best plane through the five almost coplanar atoms forms a dihedral angle of 89.56 (5)° with the benzene ring. The crystal packing is characterized by centrosymmetric dimers connected by pairs of N—H ... S hydrogen bonds.
The title compound, [Tl4(C4H9O)4], featuring a (Tl—O)4 cube, crystallizes with a quarter-molecule (located on a special position of site symmetry An external file that holds a picture, illustration, etc. Object name is e-66-m1621-efi1.jpg..) and a half-molecule (located on a special position of site symmetry 23.) in the asymmetric unit. The Tl—O bond distances range from 2.463 (12) to 2.506 (12) Å. All O—Tl—O bond angles are smaller than 90° whereas the Tl—O—Tl angles are wider than a rectangular angle.
Development of a Bioaerosol single particle detector (BIO IN) for the Fast Ice Nucleus CHamber FINCH

In this work we present the setup and first tests of our new BIO IN detector. This detector was constructed to classify atmospheric ice nuclei (IN) for their biological content. It is designed to be coupled to the Fast Ice Nucleus CHamber FINCH. If one particle acts as an ice nucleus, it will be at least partly covered with ice at the end of the development section of the FINCH chamber. The device combines an auto-fluorescence detector and a circular depolarization detector for simultaneous detection of biological material and discrimination between water droplets, ice crystals and non activated large aerosol particles. The excitation of biological material with UV light and analysis of auto-fluorescence is a common principle used for flow cytometry, fluorescence microscopy, spectroscopy and imaging. The detection of auto-fluorescence of airborne single particles demands some more experimental effort. However, expensive commercial sensors are available for special purposes, e.g. size distribution measurements. But these sensors will not fit the specifications needed for the FINCH IN counter (e.g. high sample flow of up 10 LPM). The newly developed -low cost- BIO IN sensor uses a single high-power UV LED for the electronic excitation instead of much more expensive UV lasers. Other key advantages of the new sensor are the low weight, compact size, and the little effect on the aerosol sample, which allows it to be coupled with other instruments for further analysis. The instrument will be flown on one of the first missions of the new German research aircraft "HALO" (High Altitude and LOng range).
"You've heard of Oxford Circus, you've heard of Piccadilly Circus, and this is the Rolling Stones Rock and Roll Circus" - als Rolling-Stones-Frontmann Mick Jagger diesen Satz am 11. Dezember 1968 gegen 14 Uhr in der Montur eines Zirkusdirektors in die Kameras der BBC spricht, ahnt er mit Sicherheit noch nicht, was insbesondere er in den folgenden Wochen für einen beispiellosen Zirkus rund um diese Fernsehaufzeichnung veranstalten wird. Zu diesem Zeitpunkt, da die Bänder für die Aufzeichnung des 'Rolling Stones Rock and Roll Circus' gestartet werden, glaubt Jagger wohl noch, dass die TV-Sendung ein entspanntes Aufeinandertreffen mit befreundeten Rock'n'Roll-Größen wird; er glaubt wohl noch, dass der Auftritt der Rolling Stones am Ende des Konzertes zum furiosen Abschluss wird und die Performances der übrigen Bands in den Schatten stellt; und vor allem glaubt er wohl noch, dass der 'Rock and Roll Circus' wenige Wochen nach der Aufzeichnung über die heimischen Fernseher Großbritanniens flimmert. 15 Stunden später denkt Jagger über all dies anders. Aber der Reihe nach.
Die US-amerikanische Alternative Rockband The Eels wurde 1995 durch den Songwriter Mark Oliver Everett gegründet, der der programmatische Kopf der Gruppe geblieben ist. Sie zeichnete sich von Beginn an durch eine ungewöhnliche Experimentierfreude aus, darum bemüht, immer neue unterschiedliche Klangspektren in der Musik der Band zu entfalten. Everett ist der einzige, der noch aus der Erstformation stammt; sämtliche anderen Mitglieder der Gruppe sind z.T. mehrfach ausgetauscht wurden.
Das Exil war sein Schicksal. Davon ist er zeit seines Lebens nicht losgekommen. Hans Sahl, aus Dresden gebürtig, gehörte zu jenen brillanten Intellektuellen, die bereits im geistigen Leben der Weimarer Republik eine Rolle spielten, vor den Nazis fliehen mußten und als ungeliebte Heimkehrer in der Bundesrepublik lange vergessen blieben. Erst in seinen letzten Lebensjahren, die der fast erblindete Autor in Tübingen verbrachte, wurde er mit Preisen überschüttet, wurden seine Gedichte und Romane neuaufgelegt. “Wir sind die Letzten“, hieß eine Gedichtsammlung, die 1941 erschien, in der sich dieser „Trödler des Unbegreiflichen“, wie er sich nannte, als Moralist und Wahrheitsfanatiker von hohen Graden zu erkennen gab.
Although autism spectrum disorders (ASDs) have a substantial genetic basis, most of the known genetic risk has been traced to rare variants, principally copy number variants (CNVs). To identify common risk variation, the Autism Genome Project (AGP) Consortium genotyped 1558 rigorously defined ASD families for 1 million single-nucleotide polymorphisms (SNPs) and analyzed these SNP genotypes for association with ASD. In one of four primary association analyses, the association signal for marker rs4141463, located within MACROD2, crossed the genome-wide association significance threshold of P < 5 × 10−8. When a smaller replication sample was analyzed, the risk allele at rs4141463 was again over-transmitted; yet, consistent with the winner's curse, its effect size in the replication sample was much smaller; and, for the combined samples, the association signal barely fell below the P < 5 × 10−8 threshold. Exploratory analyses of phenotypic subtypes yielded no significant associations after correction for multiple testing. They did, however, yield strong signals within several genes, KIAA0564, PLD5, POU6F2, ST8SIA2 and TAF1C.

Background: The apoptosis-inducing serine protease granzyme B (GrB) is an important factor contributing to lysis of target cells by cytotoxic lymphocytes. Expression of enzymatically active GrB in recombinant form is a prerequisite for functional analysis and application of GrB for therapeutic purposes. Methods and Findings: We investigated the influence of bacterial maltose-binding protein (MBP) fused to GrB via a synthetic furin recognition motif on the expression of the MBP fusion protein also containing an N-terminal alpha-factor signal peptide in the yeast Pichia pastoris. MBP markedly enhanced the amount of GrB secreted into culture supernatant, which was not the case when GrB was fused to GST. MBP-GrB fusion protein was cleaved during secretion by an endogenous furin-like proteolytic activity in vivo, liberating enzymatically active GrB without the need of subsequent in vitro processing. Similar results were obtained upon expression of a recombinant fragment of the ErbB2/HER2 receptor protein or GST as MBP fusions. Conclusions: Our results demonstrate that combination of MBP as a solubility enhancer with specific in vivo cleavage augments secretion of processed and functionally active proteins from yeast. This strategy may be generally applicable to improve folding and increase yields of recombinant proteins.
Background: Early inner ear development requires the strict regulation of cell proliferation, survival, migration and differentiation, coordinated by the concerted action of extrinsic and intrinsic factors. Deregulation of these processes is associated with embryonic malformations and deafness. We have shown that insulin-like growth factor I (IGF-I) plays a key role in embryonic and postnatal otic development by triggering the activation of intracellular lipid and protein kinases. RAF kinases are serine/threonine kinases that regulate the highly conserved RAS-RAF-MEK-ERK signaling cascade involved in transducing the signals from extracellular growth factors to the nucleus. However, the regulation of RAF kinase activity by growth factors during development is complex and still not fully understood.
Methodology/Principal Findings: By using a combination of qRT-PCR, Western blotting, immunohistochemistry and in situ hybridization, we show that C-RAF and B-RAF are expressed during the early development of the chicken inner ear in specific spatiotemporal patterns. Moreover, later in development B-RAF expression is associated to hair cells in the sensory patches. Experiments in ex vivo cultures of otic vesicle explants demonstrate that the influence of IGF-I on proliferation but not survival depends on RAF kinase activating the MEK-ERK phosphorylation cascade. With the specific RAF inhibitor Sorafenib, we show that blocking RAF activity in organotypic cultures increases apoptosis and diminishes the rate of cell proliferation in the otic epithelia, as well as severely impairing neurogenesis of the acoustic-vestibular ganglion (AVG) and neuron maturation.
Conclusions/Significance: We conclude that RAF kinase activity is essential to establish the balance between cell proliferation and death in neuroepithelial otic precursors, and for otic neuron differentiation and axonal growth at the AVG.
Background: Falciparum Malaria, an infectious disease caused by the apicomplexan parasite Plasmodium falciparum, is among the leading causes of death and morbidity attributable to infectious diseases worldwide. In Gabon, Central Africa, one out of four inpatients have severe malarial anemia (SMA), a life-threatening complication if left untreated. Emerging drug resistant parasites might aggravate the situation. This case control study investigates biomarkers of enhanced hemolysis in hospitalized children with either SMA or mild malaria (MM). Methods and Findings: Ninety-one children were included, thereof 39 SMA patients. Strict inclusion criteria were chosen to exclude other causes of anemia. At diagnosis, erythrophagocytosis (a direct marker for extravascular hemolysis, EVH) was enhanced in SMA compared to MM patients (5.0 arbitrary units (AU) (interquartile range (IR): 2.2–9.6) vs. 2.1 AU (IR: 1.3–3.9), p<0.01). Furthermore, indirect markers for EVH, (i.e. serum neopterin levels, spleen size enlargement and monocyte pigment) were significantly increased in SMA patients. Markers for erythrocyte ageing, such as CD35 (complement receptor 1), CD55 (decay acceleration factor) and phosphatidylserine exposure (annexin-V-binding) were investigated by flow cytometry. In SMA patients, levels of CD35 and CD55 on the red blood cell surface were decreased and erythrocyte removal markers were increased when compared to MM or reconvalescent patients. Additionally, intravascular hemolysis (IVH) was quantified using several indirect markers (LDH, alpha-HBDH, haptoglobin and hemopexin), which all showed elevated IVH in SMA. The presence of both IVH and EVH predicted the need for blood transfusion during antimalarial treatment (odds ratio 61.5, 95% confidence interval (CI): 8.9–427). Interestingly, this subpopulation is characterized by a significantly lowered reticulocyte production index (RPI, p<0.05). Conclusions: Our results show the multifactorial pathophysiology of SMA, whereby EVH and IVH play a particularly important role. We propose a model where removal of infected and non-infected erythrocytes of all ages (including reticulocytes) by EVH and IVH is a main mechanism of SMA. Further studies are underway to investigate the mechanism and extent of reticulocyte removal to identify possible interventions to reduce the risk of SMA development.
Recombinase-mediated cassette exchange (RMCE) exploits the possibility to unidirectionally exchange any genetic material flanked by heterotypic recombinase recognition sites (RRS) with target sites in the genome. Due to a limited number of available pre-fabricated target sites, RMCE in mouse embryonic stem (ES) cells has not been tapped to its full potential to date. Here, we introduce a universal system, which allows the targeted insertion of any given transcriptional unit into 85 742 previously annotated retroviral conditional gene trap insertions, representing 7013 independent genes in mouse ES cells, by RMCE. This system can be used to express any given cDNA under the control of endogenous trapped promoters in vivo, as well as for the generation of transposon ‘launch pads’ for chromosomal region-specific ‘Sleeping Beauty’ insertional mutagenesis. Moreover, transcription of the gene-of-interest is only activated upon Cre-recombinase activity, a feature that adds conditionality to this expression system, which is demonstrated in vivo. The use of the RMCE system presented in this work requires one single-cloning step followed by one overnight gateway clonase reaction and subsequent cassette exchange in ES cells with efficiencies of 40% in average.
We solved the crystal structure of a novel type of c-ring isolated from Bacillus pseudofirmus OF4 at 2.5 Å, revealing a cylinder with a tridecameric stoichiometry, a central pore, and an overall shape that is distinct from those reported thus far. Within the groove of two neighboring c-subunits, the conserved glutamate of the outer helix shares the proton with a bound water molecule which itself is coordinated by three other amino acids of outer helices. Although none of the inner helices contributes to ion binding and the glutamate has no other hydrogen bonding partner than the water oxygen, the site remains in a stable, ion-locked conformation that represents the functional state present at the c-ring/membrane interface during rotation. This structure reveals a new, third type of ion coordination in ATP synthases. It appears in the ion binding site of an alkaliphile in which it represents a finely tuned adaptation of the proton affinity during the reaction cycle. The thermodynamics of base pairing is of fundamental importance. Fluorinated base analogs are valuable tools for investigating pairing interactions. To understand the influence of direct base–base interactions in relation to the role of water, pairing free energies between natural nucleobases and fluorinated analogs are estimated by potential of mean force calculations. Compared to pairing of AU and GC, pairing involving fluorinated analogs is unfavorable by 0.5–1.0 kcal mol -1. Decomposing the pairing free energies into enthalpic and entropic contributions reveals fundamental differences for Watson–Crick pairs compared to pairs involving fluorinated analogs. These differences originate from direct base–base interactions and contributions of water. Pairing free energies of fluorinated base analogs with natural bases are less unfavorable by 0.5–1.0 kcal mol -1 compared to non-fluorinated analogs. This is attributed to stabilizing C–F…H–N dipolar interactions and stronger N…H–C hydrogen bonds, demonstrating direct and indirect influences of fluorine. 7-methyl-7H-purine and its 9-deaza analog (Z) have been suggested as members of a new class of non-fluorinated base analogs. Z is found to be the least destabilizing universal base in the context of RNA known to date. This is the first experimental evidence for nitrogen-containing heterocylces as bioisosteres of aromatic rings bearing fluorine atoms.

Rhinoliths are mineralised foreign bodies in the nasal cavity that are a chance finding at anterior rhinoscopy. Undiscovered, they grow appreciably in size and can cause a foul-smelling nasal discharge and breathing problems. Giant nasal stones are now a very rare occurrence, since improved diagnostic techniques, such as endoscopic/microscopic rhinoscopy, now make it possible to identify foreign bodies at an early stage of development. We report the case of a 37-year-old patient who, at the age of 5-6 years, introduced a foreign body, probably a stone, into his right nasal cavity. On presentation, he complained of difficulty in breathing through the right nostril that had persisted for the last 10 years. For the past four years a strong fetid smell from the nose had been apparent to those in his vicinity. Under general anaesthesia, the stone was removed in toto from the right nasal cavity. The possible genesis of the rhinolith is discussed, our case compared with those described in the literature, and possible differential diagnoses are considered.
Peroxisome proliferator-activated receptor γ (PPARγ) gained considerable interest as a therapeutic target during chronic inflammatory diseases. Remarkably, the pathogenesis of diseases such as multiple sclerosis or Alzheimer is associated with impaired PPARγ expression. Considering that regulation of PPARγ expression during inflammation is largely unknown, we were interested in elucidating underlying mechanisms. To this end, we initiated an inflammatory response by exposing primary human macrophages to lipopolysaccharide (LPS) and observed a rapid decline of PPARγ1 expression. Because promoter activities were not affected by LPS, we focused on mRNA stability and noticed a decreased mRNA half-life. As RNA stability is often regulated via 3′-untranslated regions (UTRs), we analyzed the impact of the PPARγ-3′-UTR by reporter assays using specific constructs. LPS significantly reduced luciferase activity of the pGL3-PPARγ-3′-UTR, suggesting that PPARγ1 mRNA is destabilized. Deletion or mutation of a potential microRNA-27a/b (miR-27a/b) binding site within the 3′-UTR restored luciferase activity. Moreover, inhibition of miR-27b, which was induced upon LPS exposure, partially reversed PPARγ1 mRNA decay, whereas miR-27b overexpression decreased PPARγ1 mRNA content. In addition, LPS further reduced this decay. The functional relevance of miR-27b-dependent PPARγ1 decrease was proven by inhibition or overexpression of miR-27b, which affected LPS-induced expression of the pro-inflammatory cytokines tumor necrosis factor α (TNFα) and interleukin (IL)-6. We provide evidence that LPS-induced miR-27b contributes to destabilization of PPARγ1 mRNA. Understanding molecular mechanisms decreasing PPARγ might help to better appreciate inflammatory diseases.
The human immunodeficiency virus (HIV) protease inhibitor saquinavir shows anticancer activity. Although its nitric oxide-modified derivative saquinavir-NO (saq-NO) was less toxic to normal cells, it exerted stronger inhibition of B16 melanoma growth in syngeneic C57BL/6 mice than saquinavir did. Saq-NO has been shown to block proliferation, upregulate p53 expression, and promote differentiation of C6 glioma and B16 cells. The anticancer activity of substances is frequently hampered by cancer cell chemoresistance mechanisms. Therefore, we here investigated the roles of p53 and the ATP-binding cassette (ABC) transporters P-glycoprotein (P-gp), multidrug resistance-associated protein 1 (MRP1), and breast cancer resistance protein 1 (BCRP1) in cancer cell sensitivity to saq-NO to get more information about the potential of saq-NO as anticancer drug. Saq-NO exerted anticancer effects in lower concentrations than saquinavir in a panel of human cancer cell lines. Neither p53 mutation or depletion nor expression of P-gp, MRP1, or BCRP1 affected anticancer activity of saq-NO or saquinavir. Moreover, saq-NO sensitized P-gp-, MRP1-, or BCRP1-expressing cancer cells to chemotherapy. Saq-NO induced enhanced sensitization of P-gp- or MRP1-expressing cancer cells to chemotherapy compared with saquinavir, whereas both substances similarly sensitized BCRP1-expressing cells. Washout kinetics and ABC transporter ATPase activities demonstrated that saq-NO is a substrate of P-gp as well as of MRP1. These data support the further investigation of saq-NO as an anticancer drug, especially in multidrug-resistant tumors.
Transcripts of NANOG and OCT4 have been recently identified in human t(4;11) leukemia and in a model system expressing both t(4;11) fusion proteins. Moreover, downstream target genes of NANOG/OCT4/SOX2 were shown to be transcriptionally activated. However, the NANOG1 gene belongs to a gene family, including a gene tandem duplication (named NANOG2 or NANOGP1) and several pseudogenes (NANOGP2-P11). Thus, it was unclear which of the NANOG family members were transcribed in t(4;11) leukemia cells. 5'-RACE experiments revealed novel 5'-exons of NANOG1 and NANOG2, which could give rise to the expression of two different NANOG1 and three different NANOG2 protein variants. Moreover, a novel PCR-based method was established that allows distinguishing between transcripts deriving from NANOG1, NANOG2 and all other NANOG pseudogenes (P2–P11). By applying this method, we were able to demonstrate that human hematopoietic stem cells and different leukemic cells transcribe NANOG2. Furthermore, we functionally tested NANOG1 and NANOG2 protein variants by recombinant expression in 293 cells. These studies revealed that NANOG1 and NANOG2 protein variants are functionally equivalent and activate a regulatory circuit that activates specific stem cell genes. Therefore, we pose the hypothesis that the transcriptional activation of NANOG2 represents a ‘gain-of-stem cell function’ in acute leukemia.
Epoxyeicotrienoic acids (EETs) are cytochrome P450-dependent anti-hypertensive and anti-inflammatory derivatives of arachidonic acid, which are highly abundant in the kidney and considered reno-protective. EETs are degraded by the enzyme soluble epoxide hydrolase (sEH) and sEH inhibitors are considered treatment for chronic renal failure (CRF). We determined whether sEH inhibition attenuates the progression of CRF in the 5/6-nephrectomy model (5/6-Nx) in mice. 5/6-Nx mice were treated with a placebo, an ACE-inhibitor (Ramipril, 40 mg/kg), the sEH-inhibitor cAUCB or the CYP-inhibitor fenbendazole for 8 weeks. 5/6-Nx induced hypertension, albuminuria, glomerulosclerosis and tubulo-interstitial damage and these effects were attenuated by Ramipril. In contrast, cAUCB failed to lower the blood pressure and albuminuria was more severe as compared to placebo. Plasma EET-levels were doubled in 5/6 Nx-mice as compared to sham mice receiving placebo. Renal sEH expression was attenuated in 5/6-Nx mice but cAUCB in these animals still further increased the EET-level. cAUCB also increased 5-HETE and 15-HETE, which derive from peroxidation or lipoxygenases. Similar to cAUCB, CYP450 inhibition increased HETEs and promoted albuminuria. Thus, sEH-inhibition failed to elicit protective effects in the 5/6-Nx model and showed a tendency to aggravate the disease. These effects might be consequence of a shift of arachidonic acid metabolism into the lipoxygenase pathway.
Background: A delta and C fibers are the major pain-conducting nerve fibers, activate only partly the same brain areas, and are differently involved in pain syndromes. Whether a stimulus excites predominantly A delta or C fibers is a commonly asked question in basic pain research but a quick test was lacking so far. Methodology/Principal Findings: Of 77 verbal descriptors of pain sensations, "pricking", "dull" and "pressing" distinguished best (95% cases correctly) between A delta fiber mediated (punctate pressure produced by means of von Frey hairs) and C fiber mediated (blunt pressure) pain, applied to healthy volunteers in experiment 1. The sensation was assigned to A delta fibers when "pricking" but neither "dull" nor "pressing" were chosen, and to C fibers when the sum of the selections of "dull" or "pressing" was greater than that of the selection of "pricking". In experiment 2, with an independent cohort, the three-descriptor questionnaire achieved sensitivity and specificity above 0.95 for distinguishing fiber preferential non-mechanical induced pain (laser heat, exciting A delta fibers, and 5-Hz electric stimulation, exciting C fibers). Conclusion: A three-item verbal rating test using the words "pricking", "dull", and "pressing" may provide sufficient information to characterize a pain sensation evoked by a physical stimulus as transmitted via A delta or via C fibers. It meets the criteria of a screening test by being easy to administer, taking little time, being comfortable in handling, and inexpensive while providing high specificity for relevant information.
Leukotrienes constitute a group of bioactive lipids generated by the 5-lipoxygenase (5-LO) pathway. An increasing body of evidence supports an acute role for 5-LO products already during the earliest stages of pancreatic, prostate, and colorectal carcinogenesis. Several pieces of experimental data form the basis for this hypothesis and suggest a correlation between 5-LO expression and tumor cell viability. First, several independent studies documented an overexpression of 5-LO in primary tumor cells as well as in established cancer cell lines. Second, addition of 5-LO products to cultured tumor cells also led to increased cell proliferation and activation of anti-apoptotic signaling pathways. 5-LO antisense technology approaches demonstrated impaired tumor cell growth due to reduction of 5-LO expression. Lastly, pharmacological inhibition of 5-LO potently suppressed tumor cell growth by inducing cell cycle arrest and triggering cell death via the intrinsic apoptotic pathway. However, the documented strong cytotoxic off-target effects of 5-LO inhibitors, in combination with the relatively high concentrations of 5-LO products needed to achieve mitogenic effects in cell culture assays, raise concern over the assignment of the cause, and question the relationship between 5-LO products and tumorigenesis. Background: Descending inhibitory pain control contributes to the endogenous defense against chronic pain and involves noradrenergic and serotonergic systems. The clinical efficacy of antidepressants suggests that serotonin may be particularly relevant for neuropathic pain conditions. Serotonergic signaling is regulated by synthesis, metabolisms, reuptake and receptors. To address the complexity, we used inbred mouse strains, C57BL/6J, 129 Sv, DBA/2J and Balb/c, which differ in brain serotonin levels. Results: Serotonin analysis after nerve injury revealed inter-strain differences in the adaptation of descending serotonergic fibers. Upregulation of spinal cord and midbrain serotonin was apparent only in 129 Sv mice and was associated with attenuated nerve injury evoked hyperalgesia and allodynia in this strain. The increase of dorsal horn serotonin was blocked by hemisectioning of descending fibers but not by rhizotomy of primary afferents indicating a midbrain source. Para-chlorophenylalanine-mediated serotonin depletion in spinal cord and midbrain intensified pain hypersensitivity in the nerve injury model. In contrast, chronic inflammation of the hindpaw did not evoke equivalent changes in serotonin levels in the spinal cord and midbrain and nociceptive thresholds dropped in a parallel manner in all strains. Conclusion: The results suggest that chronic nerve injury evoked hypernociception may be contributed by genetic differences of descending serotonergic inhibitory control.
Background: R-flurbiprofen, one of the enantiomers of flurbiprofen racemate, is inactive with respect to cyclooxygenase inhibition, but shows analgesic properties without relevant toxicity. Its mode of action is still unclear. Methodology/Principal Findings: We show that R-flurbiprofen reduces glutamate release in the dorsal horn of the spinal cord evoked by sciatic nerve injury and thereby alleviates pain in sciatic nerve injury models of neuropathic pain in rats and mice. This is mediated by restoring the balance of endocannabinoids (eCB), which is disturbed following peripheral nerve injury in the DRGs, spinal cord and forebrain. The imbalance results from transcriptional adaptations of fatty acid amide hydrolase (FAAH) and NAPE-phospholipase D, i.e. the major enzymes involved in anandamide metabolism and synthesis, respectively. R-flurbiprofen inhibits FAAH activity and normalizes NAPE-PLD expression. As a consequence, R-Flurbiprofen improves endogenous cannabinoid mediated effects, indicated by the reduction of glutamate release, increased activity of the anti-inflammatory transcription factor PPAR gamma and attenuation of microglia activation. Antinociceptive effects are lost by combined inhibition of CB1 and CB2 receptors and partially abolished in CB1 receptor deficient mice. R-flurbiprofen does however not cause changes of core body temperature which is a typical indicator of central effects of cannabinoid-1 receptor agonists. Conclusion: Our results suggest that R-flurbiprofen improves the endogenous mechanisms to regain stability after axonal injury and to fend off chronic neuropathic pain by modulating the endocannabinoid system and thus constitutes an attractive, novel therapeutic agent in the treatment of chronic, intractable pain.
In the past, the genetically diabetic-obese diabetes/diabetes (db/db) and obese/obese (ob/ob) mouse strains were used to investigate mechanisms of diabetes-impaired wound healing. Here we determined patterns of skin repair in genetically normal C57Bl/6J mice that were fed using a high fat diet (HFD) to induce a diabetes-obesity syndrome. Wound closure was markedly delayed in HFD-fed mice compared to mice which had received a standard chow diet (CD). Impaired wound tissue of HFD mice showed a marked prolongation of wound inflammation. Expression of vascular endothelial growth factor (VEGF) was delayed and associated with the disturbed formation of wound margin epithelia and an impaired angiogenesis in the reduced granulation tissue. Normal wound contraction was retarded and disordered. Wound disorders in obese C57Bl/6J mice were paralleled by a prominent degradation of the inhibitor of NFκB (IκB-α) in the absence of an Akt activation. By contrast to impaired wound conditions in ob/ob mice, late wounds of HFD mice did not develop a chronic inflammatory state and were epithelialized after 11 days of repair. Thus, only genetically obese and diabetic ob/ob mice finally developed chronic wounds and therefore represent a better suited experimental model to investigate diabetes-induced wound healing disorders.
This paper introduces a new methodology for the fabrication of strain-sensor elements for MEMS and NEMS applications based on the tunneling effect in nano-granular metals. The strain-sensor elements are prepared by the maskless lithography technique of focused electron-beam-induced deposition (FEBID) employing the precursor trimethylmethylcyclopentadienyl platinum [MeCpPt(Me)3]. We use a cantilever-based deflection technique to determine the sensitivity (gauge factor) of the sensor element. We find that its sensitivity depends on the electrical conductivity and can be continuously tuned, either by the thickness of the deposit or by electron-beam irradiation leading to a distinct maximum in the sensitivity. This maximum finds a theoretical rationale in recent advances in the understanding of electronic charge transport in nano-granular metals.
Tubular carbonate concretions of up to 1 m in length and perpendicular to bedding, occur abundantly in the Upper Pliensbachian (upper Amaltheus margaritatus Zone, Gibbosus Subzone) in outcrops (Fontaneilles section) in the vicinity of Rivière-sûr-Tarn, southern France. Stable isotope analyses of these concretions show negative δ13C values that decrease from the rim to the center from −18.8‰ to −25.7‰ (V-PDB), but normal marine δ18O values (−1.8‰). Carbon isotope analyses of Late Pliensbachian bulk carbonate (matrix) samples from the Fontaneilles section show clearly decreasing C-isotope values across the A. margaritatus Zone, from +1‰ to −3‰ (V-PDB). Isotope analyses of coeval belemnite rostra do not document such a negative C-isotope trend with values remaining stable around +2‰ (V-PDB). Computer tomographic (CT) scanning of the tubular concretions show multiple canals that are lined or filled entirely with pyrite. Previously, the formation of these concretions with one, two, or more central tubes, has been ascribed to the activity of an enigmatic organism, possibly with annelid or arthropod affinities, known as Tisoa siphonalis. Our results suggest tisoan structures are abiogenic. Based on our geochemical analyses and sedimentological observations we suggest that these concretions formed as a combination of the anaerobic oxidation of methane (AOM) and sulfate reduction within the sediment. Fluids rich in methane and/or hydrocarbons likely altered local bulk rock carbon isotope records, but did not affect the global carbon cycle. Interestingly, Tisoa siphonalis has been described from many locations in the Grands Causses Basin in southern France, and from northern France and Luxemburg, always occurring at the same stratigraphic level. Upper Pliensbachian authigenic carbonates thus possibly cover an area of many thousand square kilometers. Greatly reduced sedimentation rates are needed to explain the stabilization of the sulfate-methane transition zone in the sedimentary column in order for the tubular concretions to form. Late Pliensbachian cooling, reducing run-off, and/or the influx of colder water and more vigorous circulation could be responsible for a halt in sedimentation. At the same time (thermogenic) methane may have destabilized during a major phase of Late Pliensbachian sea level fall. As such Tisoa siphonalis is more than a geological curiosity, and its further study could prove pivotal in understanding Early Jurassic paleoenvironmental change.

The continuous progress in the structural and functional characterization of aquaporins increasingly attracts attention to study their roles in certain mammalian diseases. Although several structures of aquaporins have already been solved by crystallization, the challenge of producing sufficient amounts of functional proteins still remains. CF (cell free) expression has emerged in recent times as a promising alternative option in order to synthesize large quantities of membrane proteins, and the focus of this report was to evaluate the potential of this technique for the production of eukaryotic aquaporins. We have selected the mouse aquaporin 4 as a representative of mammalian aquaporins. The protein was synthesized in an E. coli extract based cell-free system with two different expression modes, and the efficiencies of two modes were compared. In both, the P-CF (cell-free membrane protein expression as precipitate) mode generating initial aquaporin precipitates as well as in the D-CF (cell-free membrane protein expression in presence of detergent) mode, generating directly detergent solubilized samples, we were able to obtain mg amounts of protein per ml of cell-free reaction. Purified aquaporin samples solubilized in different detergents were reconstituted into liposomes, and analyzed for the water channel activity. The calculated Pf value of proteoliposome samples isolated from the D-CF mode was 133 µm/s at 10°C, which was 5 times higher as that of the control. A reversible inhibitory effect of mercury chloride was observed, which is consistent with previous observations of in vitro reconstituted aquaporin 4. In this study, a fast and convenient protocol was established for functional expression of aquaporins, which could serve as basis for further applications such as water filtration.
Proteins of the Omp85 family are conserved in all kingdoms of life. They mediate protein transport across or protein insertion into membranes and reside in the outer membranes of Gram-negative bacteria, mitochondria, and chloroplasts. Omp85 proteins contain a C-terminal transmembrane β-barrel and a soluble N terminus with a varying number of polypeptide-transport-associated or POTRA domains. Here we investigate Omp85 from the cyanobacterium Anabaena sp. PCC 7120. The crystallographic three-dimensional structure of the N-terminal region shows three POTRA domains, here named P1 to P3 from the N terminus. Molecular dynamics simulations revealed a hinge between P1 and P2 but in contrast show that P2 and P3 are fixed in orientation. The P2-P3 arrangement is identical as seen for the POTRA domains from proteobacterial FhaC, suggesting this orientation is a conserved feature. Furthermore, we define interfaces for protein-protein interaction in P1 and P2. P3 possesses an extended loop unique to cyanobacteria and plantae, which influences pore properties as shown by deletion. It now becomes clear how variations in structure of individual POTRA domains, as well as the different number of POTRA domains with both rigid and flexible connections make the N termini of Omp85 proteins versatile adaptors for a plentitude of functions.
Moderately elevated levels of plasma plant sterols have been suspected to be causally involved in atherosclerosis. The aim of this study was to investigate whether plant sterols and other markers of sterol metabolism predicted all-cause and cardiovascular mortality in participants of the Ludwigshafen Risk and Cardiovascular health (LURIC) study. A total of 1,257 individuals who did not use statins and at baseline had a mean (± SD) age of 62.8 (± 11.0) years were included in the present analysis. Lathosterol, cholestanol, campesterol, and sitosterol were measured to estimate cholesterol synthesis and absorption. The mean (± SD) time of the follow-up for all-cause and cardiovascular mortality was 7.32 (± 2.3) years. All-cause (P = 0.001) and cardiovascular (P = 0.006) mortality were decreased in the highest versus the lowest lathosterol to cholesterol tertile. In contrast, subjects in the third cholestanol to cholesterol tertile had increased all-cause (P < 0.001) and cardiovascular mortality (P = 0.010) compared with individuals in the first tertile. The third campesterol to cholesterol tertile was associated with increased all-cause mortality (P = 0.025). Sitosterol to cholesterol tertiles were not significantly related to all-cause or cardiovascular mortality. The data suggest that high absorption and low synthesis of cholesterol predict increased all-cause and cardiovascular mortality in LURIC participants.
Surface measurements of aerosol and ice nuclei (IN) at a Central European mountain site during an episode of dust transport from the Sahara are presented. Ice nuclei were sampled by electrostatic precipitation on silicon wafers and were analyzed in an isothermal static vapor diffusion chamber. The transport of mineral dust is simulated by the Eulerian regional dust model DREAM. Ice nuclei and mineral dust are significantly correlated, in particular IN number concentration and aerosol surface area. The ice nucleating characteristics of the aerosol as analyzed with respect to temperature and supersaturation are similar during the dust episode than during the course of the year. This suggests that dust may be a main constituent of ice nucleating aerosols in Central Europe.

Transport is simulated by the Eulerian regional dust model DREAM. Ice nuclei and mineral dust are significantly correlated. The highest correlation is found between IN concentration and aerosol surface area. The ice nucleating characteristics of the aerosol with respect to temperature and supersaturation are similar during the dust episode than during the course of the year. This suggests that dust is always a dominant constituent of ice nucleating aerosols in Central Europe.
Background: Many disabling human retinal disorders involve the central retina, particularly the macula. However, the commonly used rodent models in research, mouse and rat, do not possess a macula. The purpose of this study was to identify small laboratory rodents with a significant central region as potential new models for macular research.
Methodology/Principal Findings: Gerbillus perpallidus, Meriones unguiculatus and Phodopus campbelli, laboratory rodents less commonly used in retinal research, were subjected to confocal scanning laser ophthalmoscopy (cSLO), fluorescein and indocyanine green angiography, and spectral-domain optical coherence tomography (SD-OCT) using standard equipment (Heidelberg Engineering HRA1 and Spectralis™) adapted to small rodent eyes. The existence of a visual streak-like pattern was assessed on the basis of vascular topography, retinal thickness, and the topography of retinal ganglion cells and cone photoreceptors. All three species examined showed evidence of a significant horizontal streak-like specialization. cSLO angiography and retinal wholemounts revealed that superficial retinal blood vessels typically ramify and narrow into a sparse capillary net at the border of the respective area located dorsal to the optic nerve. Similar to the macular region, there was an absence of larger blood vessels in the streak region. Furthermore, the thickness of the photoreceptor layer and the population density of neurons in the ganglion cell layer were markedly increased in the visual streak region.
Conclusions/Significance: The retinal specializations of Gerbillus perpallidus, Meriones unguiculatus and Phodopus campbelli resemble features of the primate macula. Hence, the rodents reported here may serve to study aspects of macular development and diseases like age-related macular degeneration and diabetic macular edema, and the preclinical assessment of therapeutic strategies.

Entsprechend kann wIRA sehr gut zur Verbesserung der Wundheilung bei akuten und chronischen Wunden eingesetzt werden.
Poster presentation at 5th German Conference on Cheminformatics: 23. CIC-Workshop Goslar, Germany. 8-10 November 2009 We demonstrate the theoretical and practical application of modern kernel-based machine learning methods to ligand-based virtual screening by successful prospective screening for novel agonists of the peroxisome proliferator-activated receptor gamma (PPARgamma) [1]. PPARgamma is a nuclear receptor involved in lipid and glucose metabolism, and related to type-2 diabetes and dyslipidemia. Applied methods included a graph kernel designed for molecular similarity analysis [2], kernel principle component analysis [3], multiple kernel learning [4], and, Gaussian process regression [5]. In the machine learning approach to ligand-based virtual screening, one uses the similarity principle [6] to identify potentially active compounds based on their similarity to known reference ligands. Kernel-based machine learning [7] uses the "kernel trick", a systematic approach to the derivation of non-linear versions of linear algorithms like separating hyperplanes and regression. Prerequisites for kernel learning are similarity measures with the mathematical property of positive semidefiniteness (kernels). The iterative similarity optimal assignment graph kernel (ISOAK) [2] is defined directly on the annotated structure graph, and was designed specifically for the comparison of small molecules. In our virtual screening study, its use improved results, e.g., in principle component analysis-based visualization and Gaussian process regression. Following a thorough retrospective validation using a data set of 176 published PPARgamma agonists [8], we screened a vendor library for novel agonists. Subsequent testing of 15 compounds in a cell-based transactivation assay [9] yielded four active compounds. The most interesting hit, a natural product derivative with cyclobutane scaffold, is a full selective PPARgamma agonist (EC50 = 10 ± 0.2 microM, inactive on PPARalpha and PPARbeta/delta at 10 microM). We demonstrate how the interplay of several modern kernel-based machine learning approaches can successfully improve ligand-based virtual screening results.
Influenza A (H1N1) 2009 : impact on Frankfurt in due consideration of health care and public health

Background: In April 2009 a novel influenza A H1N1/2009 virus was identified in Mexico and in the United States which quickly spread around the world. Most of the countries established infection surveillance systems in order to track the number of (laboratory-confirmed) H1N1 cases, hospitalizations and deaths. Methods: The impact of the emergence of the novel pandemic (H1N1) 2009 virus on Frankfurt was statistically evaluated by the Health Protection Authority, City of Frankfurt am Main. Vaccination rates of the health care workers (HCWs) of the University Hospital Frankfurt were measured by the Occupational Health Service. Results: Although the virulence of pandemic (H1N1) 2009 seems to be comparable with seasonal influenza, a major patient load and wave of hospital admissions occurred in the summer of 2009. Even though the 2009 vaccination rate of the University Hospital Frankfurt (seasonal influenza [40.5%], swine flu [36.3%]) is better than the average annual uptake of influenza vaccine in the German health care system (approximately 22% for seasonal and 15% for swine flu), vaccination levels remain insufficient. However, physicians were significantly (p < 0.001) more likely to have been vaccinated against swine flu and seasonal influenza than nurses. Conclusions: The outbreak of the pandemic (H1N1) 2009 in April 2009 provided a major challenge to health services around the world. Nosocomial transmission of H1N1/2009 has been documented. Present experience should be used to improve pandemic preparedness plans and vaccination programs ought to target as many HCWs as possible.

Products of ene reactions of isobutene and tBu2Si=N-SiRtBu2 (R = tBu, Ph) are formed. X-Ray quality crystals of H2C=C(CH2SitBu2-NH-SiPhtBu2)2 (monoclinic, space group C2/c, Z = 4) were grown from a benzene solution at ambient temperature, whereas single crystals of H2C=C(CH2SitBu2-NH-SitBu3)2 (monoclinic, space group P21, Z = 2) were obtained by recrystallization from THF.

If insufficiently treated, Lyme borreliosis can evolve into an inflammatory disorder affecting skin, joints, and the CNS. Early innate immunity may determine host responses targeting infection. Thus, we sought to characterize the immediate cytokine storm associated with exposure of PBMC to moderate levels of live Borrelia burgdorferi. Since Th17 cytokines are connected to host defense against extracellular bacteria, we focused on interleukin (IL)-17 and IL-22. Here, we report that, despite induction of inflammatory cytokines including IL-23, IL-17 remained barely detectable in response to B. burgdorferi. In contrast, T cell-dependent expression of IL-22 became evident within 10 h of exposure to the spirochetes. This dichotomy was unrelated to interferon-gamma but to a large part dependent on caspase-1 and IL-1 bioactivity derived from monocytes. In fact, IL-1beta as a single stimulus induced IL-22 but not IL-17. Neutrophils display antibacterial activity against B. burgdorferi, particularly when opsonized by antibodies. Since neutrophilic inflammation, indicative of IL-17 bioactivity, is scarcely observed in Erythema migrans, a manifestation of skin inflammation after infection, protective and antibacterial properties of IL-22 may close this gap and serve essential functions in the initial phase of spirochete infection.
Background In October 2007, the working group CEN/TC 216 of the European Committee for standardisation suggested that the Sabin oral poliovirus vaccine type 1 strain (LSc-2ab) presently used for virucidal tests should be replaced by another attenuated vaccine poliovirus type 1 strain, CHAT. Both strains were historically used as oral vaccines, but the Sabin type 1 strain was acknowledged to be more attenuated. In Germany, vaccination against poliomyelitis was introduced in 1962 using the oral polio vaccine (OPV) containing Sabin strain LSc-2ab. The vaccination schedule was changed from OPV to an inactivated polio vaccine (IPV) containing wild polio virus type 1 strain Mahoney in 1998. In the present study, we assessed potential differences in neutralising antibody titres to Sabin and CHAT in persons with a history of either OPV, IPV, or OPV with IPV booster. Methods Neutralisation poliovirus antibodies against CHAT and Sabin 1 were measured in sera of 41 adults vaccinated with OPV. Additionally, sera from 28 children less than 10 years of age and immunised with IPV only were analysed. The neutralisation assay against poliovirus was performed according to WHO guidelines. Results The neutralisation activity against CHAT in adults with a complete OPV vaccination series was significantly lower than against Sabin poliovirus type 1 strains (Wilcoxon signed-rank test P < 0.025). In eight sera, the antibody titres measured against CHAT were less than 8, although the titre against Sabin 1 varied between 8 and 64. Following IPV booster, anti-CHAT antibodies increased rapidly in sera of CHAT-negative adults with OPV history. Sera from children with IPV history neutralised CHAT and Sabin 1 strains equally. Conclusion The lack of neutralising antibodies against the CHAT strain in persons vaccinated with OPV might be associated with an increased risk of reinfection with the CHAT polio virus type 1, and this implies a putative risk of transmission of the virus to polio-free communities. We strongly suggest that laboratory workers who were immunised with OPV receive a booster vaccination with IPV before handling CHAT in the laboratory.
