The large conductance voltage- and Ca2+-activated potassium (BK) channel has been suggested to play an important role in the signal transduction process of cochlear inner hair cells. BK channels have been shown to be composed of the pore-forming alpha-subunit coexpressed with the auxiliary beta-1-subunit. Analyzing the hearing function and cochlear phenotype of BK channel alpha-(BKalpha–/–) and beta-1-subunit (BKbeta-1–/–) knockout mice, we demonstrate normal hearing function and cochlear structure of BKbeta-1–/– mice. During the first 4 postnatal weeks also, BKalpha–/– mice most surprisingly did not show any obvious hearing deficits. High-frequency hearing loss developed in BKalpha–/– mice only from ca. 8 weeks postnatally onward and was accompanied by a lack of distortion product otoacoustic emissions, suggesting outer hair cell (OHC) dysfunction. Hearing loss was linked to a loss of the KCNQ4 potassium channel in membranes of OHCs in the basal and midbasal cochlear turn, preceding hair cell degeneration and leading to a similar phenotype as elicited by pharmacologic blockade of KCNQ4 channels. Although the actual link between BK gene deletion, loss of KCNQ4 in OHCs, and OHC degeneration requires further investigation, data already suggest human BK-coding slo1 gene mutation as a susceptibility factor for progressive deafness, similar to KCNQ4 potassium channel mutations. © 2004, The National Academy of Sciences. Oxidative stress attenuates the NO-cGMP pathway, e.g. in the vascular system, through scavenging of free NO radicals by superoxide O2•-, by inactivation of soluble guanylyl cyclase (sGC) via oxidation of its central Fe2+ ion, and by down-regulation of sGC protein levels. While the former pathways are well established, the molecular mechanisms underlying the latter are still obscure. Using oxidative sGC inhibitor ODQ we demonstrate rapid down-regulation of sGC protein in mammalian cells. Co-incubation with proteasomal inhibitor MG132 results in accumulation of ubiquitinated sGC whereas sGC activator BAY 58–2667 prevents ubiquitination. ODQ-induced down-regulation of sGC is mediated through selective ubiquitination of its b subunit, and BAY 58–2667 abrogates this effect. Ubiquitination of sGC-b is dramatically enhanced by E3 ligase CHIP. Our data indicate that oxidative stress promotes ubiquitination of sGC b subunit through E3 ligase CHIP, and that sGC activator 58–2667 reverts this effect, most likely through stabilization of the heme-free b subunit. Thus the deleterious effects of oxidative stress can be counter-balanced by an activator of a key enzyme of vascular homeostasis.
Zwischen Macht und Gerechtigkeit : Zustand und Perspektiven des nuklearen Nichtverbreitungsregimes

Das nukleare Nichtverbreitungsregime weist eine erstaunliche Erfolgsstory auf. Die Zahl der Staaten, die kernwaffenorientierte Aktivitäten in verschiedenen Stadien des Fortschritts aufgegeben haben, ist weitaus größer als die der Kernwaffenstaaten. Auf dem Höhepunkt seines Erfolgs droht dem Regime jedoch Ungemach aus zwei Richtungen: Eine kleine Anzahl von „Ausbrechern“ und „Außenseitern“ unterminiert seine zentrale Zielsetzung, die Weiterverbreitung dieser Waffen aufzuhalten. Die Verweigerungshaltung der Kernwaffenstaaten gegenüber ihrer Abrüstungspflicht verstößt gegen den im Regime eingelassenen Gerechtigkeitsgrundsatz und zerstört damit seine Legitimität. Diese Erkenntnis ist mittlerweile aus dem „Ghetto“ der unverdrossenen Abrüstungsenthusiasten in den sicherheitspolitischen Mainstream vorgedrungen. Von ihrer Durchsetzungsfähigkeit wird letztlich die Zukunft des Regimes abhängen – mit weitreichenden Konsequenzen für die globale Sicherheit.
Der „demokratische Frieden“ und seine außenpolitischen Konsequenzen Demokratien führen gegeneinander keine Kriege; oder jedenfalls „fast“ keine. Dieser statistische Befund ist ziemlich robust gegenüber Veränderungen in der Definition von „Demokratie“ und von „Krieg“. Demokratien erfreuen sich überdies im Durchschnitt größeren Wohlstands, vermeiden erfolgreich Hungersnöte, bieten ihren Bürgerinnen und Bürgern mehr Freiheit und lassen sich eher auf internationale Organisationen und auf die Rechtsbindung in internationalen Verträgen ein als Staaten mit anderen Regierungsformen; dies sind natürlich Durchschnittswerte, von denen es Abweichungen gibt. Aber die Nachricht ist ziemlich klar: Demokratien bieten eine vergleichsweise bessere Form von „Good Governance“ als andere Systeme.
In diesem Aufsatz konzentriere ich mich auf einen wichtigen und überaus kühnen, praktisch-politischen Vorschlag des Projekts der kosmopolitischen Demokratie (Archibugi/Held 1995; Archibugi et al. 1998; Beck 2004; Held 1998): auf die Parlamentarisierung der Weltpolitik (Übersicht bei Bienen et al. 1998). Das kosmopolitische Projekt unterstellt, dass die Vergemeinschaftung der Menschheit einen Grad erreicht hat, der es erlaubt oder gar erfordert, die Herstellung von Recht – einschließlich von Weltbürgerrecht (Brunkhorst et al. 1999) und die Kontrolle seiner richtigen exekutiven Anwendung in die Hände einer globalen Legislative zu geben.
Background: Nitric oxide (NO) is an essential vasodilator. In vascular diseases, oxidative stress attenuates NO signaling by both chemical scavenging of free NO and oxidation and down-regulation of its major intracellular receptor, the alpha/beta heterodimeric heme-containing soluble guanylate cyclase (sGC). Oxidation can also induce loss of sGC's heme and responsiveness to NO.
Results: sGC activators such as BAY 58-2667 bind to oxidized/heme-free sGC and reactivate the enzyme to exert disease-specific vasodilation. Here we show that oxidation-induced down-regulation of sGC protein extends to isolated blood vessels. Mechanistically, degradation was triggered through sGC ubiquitination and proteasomal degradation. The heme-binding site ligand, BAY 58-2667, prevented sGC ubiquitination and stabilized both alpha and beta subunits.
Conclusion: Collectively, our data establish oxidation-ubiquitination of sGC as a modulator of NO/cGMP signaling and point to a new mechanism of action for sGC activating vasodilators by stabilizing their receptor, oxidized/heme-free sGC.
Die Regime gegen Massenvernichtungswaffen erfüllen wichtige sicherheitspolitische Funktionen und tragen dazu bei, Terrorismus mit diesen Waffen zu verhindern. Eine regimetheoretische Analyse zeigt für alle drei Regime stabilisierende und destabilisierende Tendenzen sowie eine Spaltung zwischen Nord und Süd. Im nuklearen Nichtverbreitungsregime wirkt die Diskriminierung zwischen Kernwaffen- und Nichtkernwaffenstaaten in Verbindung mit der mangelhaften Umsetzung der Abrüstungsverpfl ichtung destabilisierend. Das Biowaffen-Regime zeigt eine neue Dynamik zum Thema Biosicherheit, aber auch anhaltende Konfl ikte um Technologieaustausch und Verifikation. Obwohl das CWÜ derzeit am stabilsten erscheint, gilt es, drohende Probleme z.B. im Bereich Verifi kation und Abrüstung abzuwenden. Wenn die Regime ihrer Aufgabe effektiv nachkommen sollen, müssen die strukturellen Defizite bearbeitet sowie jeweils die Abrüstungs-, Nichtverbreitungs- und Kooperationsbestimmungen gleichermaßen vollständig und ausgewogen umgesetzt werden.
Rüstungskontrolle entwickelte sich während des Ost-West-Konflikts zu einem wichtigen Element für die Stabilisierung des internationalen Systems und zur Verhütung eines Atomkriegs, wobei jedoch die Rahmenbedingungen der bipolaren Blockkonfrontation immer wieder einschränkend wirkten. Das Ende des Kalten Krieges brachte zunächst eine Welle des Optimismus und der Hoffnung mit sich, dass nun größere Fortschritte in der Rüstungskontrolle möglich sein sollten. Und in der Tat stellten sich in der ersten Hälfte der 1990er Jahre Erfolge ein: Unter anderem wurden die Zahl der atomaren Sprengköpfe reduziert, Chemiewaffen und Antipersonenminen verboten, der nukleare Umfassende Teststoppvertrag (CTBT) abgeschlossen und der Vertrag über konventionelle Streitkräfte in Europa (KSE) an die neuen Bedingungen angepasst. Ab Mitte der 1990er Jahre setzte jedoch eine Stagnation in der Rüstungskontrolle ein, die sich spätestens mit der Amtsübernahme der Bush-Administration zu einer handfesten Krise auswuchs. Von manchen Beobachter/innen und Praktiker/innen wurde gar das Ende der Rüstungskontrolle – zumindest in ihrer bisher bekannten Form – postuliert oder diagnostiziert.
The first measurement of two-pion Bose–Einstein correlations in central Pb–Pb collisions at √sNN=2.76 TeV at the Large Hadron Collider is presented. We observe a growing trend with energy now not only for the longitudinal and the outward but also for the sideward pion source radius. The pion homogeneity volume and the decoupling time are significantly larger than those measured at RHIC.
Inclusive transverse momentum spectra of primary charged particles in Pb–Pb collisions at √sNN=2.76 TeV have been measured by the ALICE Collaboration at the LHC. The data are presented for central and peripheral collisions, corresponding to 0–5% and 70–80% of the hadronic Pb–Pb cross section. The measured charged particle spectra in |η|<0.8 and 0.3<pT<20 GeV/c are compared to the expectation in pp collisions at the same sNN, scaled by the number of underlying nucleon–nucleon collisions. The comparison is expressed in terms of the nuclear modification factor RAA. The result indicates only weak medium effects (RAA≈0.7) in peripheral collisions. In central collisions, RAA reaches a minimum of about 0.14 at pT=6–7 GeV/c and increases significantly at larger pT. The measured suppression of high-pT particles is stronger than that observed at lower collision energies, indicating that a very dense medium is formed in central Pb–Pb collisions at the LHC.
