Der Hochmoorlaufkäfer (Carabus menetriesi pacholei) ist eine prioritäre Anhang-Art der FFH-Richtlinie und Art mit hoher Schutzverantwortung Deutschlands. Seine Lebensräume liegen in noch weitgehend intakten Hoch-, Übergangs- und Hochlagen-Quellmooren einschließlich von Moorwäldern. In einem prognostizierten, wärmer und trockener werdenden Klima werden einige Vorkommen außerhalb der hier vorgelegten Klimahülle zu liegen kommen und daher für die Art voraussichtlich verloren gehen. Das gilt in besonderem Maße für die endemischen Vorkommen im Voralpengebiet. Andere Vorkommen werden nur zu halten sein, wenn jegliche Anstrengungen unternommen werden, einen naturnahen Wasserhaushalt wiederherzustellen. Die Klimahülle für die Nominatform reicht etwas weiter in den trockenen und auch warmen Bereich, erfordert aber ebenfalls sämtliche Bemühungen zu ihrem Erhalt. Zu den notwendigen Maßnahmen gehört unter anderem auch die konsequente Erhaltung naturnaher Moor- und Moorrandwälder. Eine Theorie zur Erklärung der disjunkten Verbreitung im Voralpengebiet basierend auf der Vereisungsgeschichte wird präsentiert, da die Klimaverhältnisse allein die Verbreitung nicht erklären können.

Abstract: Quer zur Standardrezeption analysiert Claudia Bruns in ihrer Dissertation Blühers Konzeption von Mann, Männlichkeit und mann-männlicher Beziehung unter besonderer Berücksichtigung damit einhergehender Ausschlüsse. Zentral legt sie das Konzept des Männerbundes zugrunde, das den Dreh- und Angelpunkt der vorliegenden Untersuchung bildet, und zieht die Theorien Blühers als paradigmatisches Beispiel dafür heran. Dieses Vorgehen ist aus verschiedenen Gründen klug gewählt und vorteilhaft. Bislang ist kaum der ideologisch vielschichtige Gehalt von Diskussionen, wie sie Blüher führte und führen konnte, in die Geschichts- und Gesellschaftswissenschaften eingegangen. Zudem gab es bislang kein Gesamtverzeichnis der Schriften Blühers, wodurch die Rezeption erschwert wurde, und das sich nun im Anhang der Arbeit dankenswerterweise findet.
Zinc finger proteins (ZNF) are a large group of transcription factors with diverse functions. We recently discovered that endothelial cells harbour a specific mechanism to limit the action of ZNF354C, whose function in endothelial cells is unknown. Given that ZNF354C has so far only been studied in bone and tumour, its function was determined in endothelial cells. ZNF354C is expressed in vascular cells and localises to the nucleus and cytoplasm. Overexpression of ZNF354C in human endothelial cells results in a marked inhibition of endothelial sprouting. RNA-sequencing of human microvascular endothelial cells with and without overexpression of ZNF354C revealed that the protein is a potent transcriptional repressor. ZNF354C contains an active KRAB domain which mediates this suppression as shown by mutagenesis analysis. ZNF354C interacts with dsDNA, TRIM28 and histones, as observed by proximity ligation and immunoprecipitation. Moreover, chromatin immunoprecipitation revealed that the ZNF binds to specific endothelial-relevant target-gene promoters. ZNF354C suppresses these genes as shown by CRISPR/Cas knockout and RNAi. Inhibition of endothelial sprouting by ZNF354C is dependent on the amino acids DV and MLE of the KRAB domain. These results demonstrate that ZNF354C is a repressive transcription factor which acts through a KRAB domain to inhibit endothelial angiogenic sprouting.
In the past, the genetically diabetic-obese diabetes/diabetes (db/db) and obese/obese (ob/ob) mouse strains were used to investigate mechanisms of diabetes-impaired wound healing. Here we determined patterns of skin repair in genetically normal C57Bl/6J mice that were fed using a high fat diet (HFD) to induce a diabetes-obesity syndrome. Wound closure was markedly delayed in HFD-fed mice compared to mice which had received a standard chow diet (CD). Impaired wound tissue of HFD mice showed a marked prolongation of wound inflammation. Expression of vascular endothelial growth factor (VEGF) was delayed and associated with the disturbed formation of wound margin epithelia and an impaired angiogenesis in the reduced granulation tissue. Normal wound contraction was retarded and disordered. Wound disorders in obese C57Bl/6J mice were paralleled by a prominent degradation of the inhibitor of NFκB (IκB-α) in the absence of an Akt activation. By contrast to impaired wound conditions in ob/ob mice, late wounds of HFD mice did not develop a chronic inflammatory state and were epithelialized after 11 days of repair. Thus, only genetically obese and diabetic ob/ob mice finally developed chronic wounds and therefore represent a better suited experimental model to investigate diabetes-induced wound healing disorders.
In der vorliegenden Arbeit wurde ein tierexperimentelles Wundheilungsmodell etabliert, welches nach Kultivierung und Transplantation autologer Keratinozyten auf einer biologische Trägermembran aus Hyaluronsäure ein Vergleich mit verschiedenen Wundauflagen im Rahmen einer standardisierten Wundbehandlung erlaubte. Acht narkotisierten Schweinen wurden paravertebral 6 Polytetraflouroethylen-Kammern implantiert und die entstandenen Vollhautwunden mit verschiedenen Wundauflagen konditioniert. Parallel dazu erfolgte die Gewinnung und Kultivierung autologer Keratinozyten auf dem Zellkulturträger Laserskin – einer Hyaluronsäureestermembran. Die Transplantation der Zellen erfolgte 1 Woche nach Wundsetzung und anschließend wurden die Wunden mit den verwendeten Wundauflagen Jelonet, Schaumstoff und Hyalofill-F, einem Hyaluronsäureesterfleece, zweitägig über einen Zeitraum von 3 Wochen behandelt. Zu fest definierten Zeitpunkten erfolgten die vergleichenden photooptischen Dokumentationen und histologischen Untersuchungen (Hämatoxilin-Eosin-Färbung, Bindegewebsfärbung nach Goldner, Elastika-Anfärbung nach Weigert) der Wunden. Zur Bestimmung der biomechanischen Narbenqualität wurde das exzidierte Narbengewebe am Versuchsende 21 Tage nach Transplantation mechanischen Zugversuchen im Laserextensiometer zugeführt und die Zugfestigkeit und Dehnung bei Maximalspannung des Gewebes bestimmt. Sowohl die mikro- und makroskopischen als auch die biomechanischen Eigenschaften der untersuchten Wunden zeigten unabhängig von dem untersuchten Tier, der untersuchten Wunde bzw. der Wundbehandlung und unabhängig vom Zeitpunkt das einheitliche Bild eines neu entstandenen Narbengewebes ohne Anzeichen einer Epithelneubildung und ohne signifikante Unterschiede hinsichtlich der Zugfestigkeit und des Dehnungsverhaltens. Verschiedene Ursachen mögen für die ausbleibende Reepithelialisierung verantwortlich sein. Es konnte jedoch mit Hilfe der hier vorgestellten Wundheilungsstudie mit verschiedenen Wundauflagen an einem Hausschweinemodell eine systematische, reproduzierbare und standardisierte Methode entwickelt werden, um Narbengewebe hinreichend zu beschreiben. Weitere Anstrengungen werden noch notwendig sein, um die Lücke zwischen tierexperimentellen Studien und bereits klinisch erfolgreich angewandten Transplantation autologer Keratinozyten beim Menschen zu schließen.
Background Since June 2002, revised regulations in Germany have required "Emergency Medical Care" as an interdisciplinary subject, and state that emergency treatment should be of increasing importance within the curriculum. A survey of the current status of undergraduate medical education in emergency medical care establishes the basis for further committee work. Methods Using a standardized questionnaire, all medical faculties in Germany were asked to answer questions concerning the structure of their curriculum, representation of disciplines, instructors' qualifications, teaching and assessment methods, as well as evaluation procedures. Results Data from 35 of the 38 medical schools in Germany were analysed. In 32 of 35 medical faculties, the local Department of Anaesthesiology is responsible for the teaching of emergency medical care; in two faculties, emergency medicine is taught mainly by the Department of Surgery and in another by Internal Medicine. Lectures, seminars and practical training units are scheduled in varying composition at 97% of the locations. Simulation technology is integrated at 60% (n=21); problem-based learning at 29% (n=10), e-learning at 3% (n=1), and internship in ambulance service is mandatory at 11% (n=4). In terms of assessment methods, multiple-choice exams (15 to 70 questions) are favoured (89%, n=31), partially supplemented by open questions (31%, n=11). Some faculties also perform single practical tests (43%, n=15), objective structured clinical examination (OSCE; 29%, n=10) or oral examinations (17%, n=6). Conclusion Emergency Medical Care in undergraduate medical education in Germany has a practical orientation, but is very inconsistently structured. The innovative options of simulation technology or state-of-the-art assessment methods are not consistently utilized. Therefore, an exchange of experiences and concepts between faculties and disciplines should be promoted to guarantee a standard level of education in emergency medical care.
Good quality data on precipitation are a prerequisite for applications like short-term weather forecasts, medium-term humanitarian assistance, and long-term climate modelling. In Sub-Saharan Africa, however, the meteorological station networks are frequently insufficient, as in the Cuvelai-Basin in Namibia and Angola. This paper analyses six rainfall products (ARC2.0, CHIRPS2.0, CRU-TS3.23, GPCCv7, PERSIANN-CDR, and TAMSAT) with respect to their performance in a crop model (APSIM) to obtain nutritional scores of a household’s requirements for dietary energy and further macronutrients. All products were calibrated to an observed time series using Quantile Mapping. The crop model output was compared against official yield data. The results show that the products (i) reproduce well the Basin’s spatial patterns, and (ii) temporally agree to station records (r = 0.84). However, differences exist in absolute annual rainfall (range: 154 mm), rainfall intensities, dry spell duration, rainy day counts, and the rainy season onset. Though calibration aligns key characteristics, the remaining differences lead to varying crop model results. While the model well reproduces official yield data using the observed rainfall time series (r = 0.52), the products’ results are heterogeneous (e.g., CHIRPS: r = 0.18). Overall, 97% of a household’s dietary energy demand is met. The study emphasizes the importance of considering the differences among multiple rainfall products when ground measurements are scarce.
We report the results of the femtoscopic analysis of pairs of identical pions measured in p-Pb collisions at sNN−−−√=5.02 TeV. Femtoscopic radii are determined as a function of event multiplicity and pair momentum in three spatial dimensions. As in the pp collision system, the analysis is complicated by the presence of sizable background correlation structures in addition to the femtoscopic signal. The radii increase with event multiplicity and decrease with pair transverse momentum. When taken at comparable multiplicity, the radii measured in p-Pb collisions, at high multiplicity and low pair transverse momentum, are 10-20% higher than those observed in pp collisions but below those observed in A-A collisions. The results are compared to hydrodynamic predictions at large event multiplicity as well as discussed in the context of calculations based on gluon saturation.
