Current research on theory and practice of digital libraries: best papers from TPDL 2019 & 2020

This volume presents a special issue on selected papers from the 2019 & 2020 editions of the International Conference on Theory and Practice of Digital Libraries (TPDL). They cover different research areas within Digital Libraries, from Ontology and Linked Data to quality in Web Archives and Topic Detection. We first provide a brief overview of both TPDL editions, and we introduce the selected papers.

This volume presents a special issue on selected papers from the 2019 & 2020 editions of the International Conference on Theory and Practice of Digital Libraries (TPDL). They cover different research areas within Digital Libraries, from Ontology and Linked Data to quality in Web Archives and Topic Detection. We first provide a brief overview of both TPDL editions, and we introduce the selected papers.
The inclusive charged particle transverse momentum distribution is measured in proton–proton collisions at s=900 GeV at the LHC using the ALICE detector. The measurement is performed in the central pseudorapidity region (|η|<0.8) over the transverse momentum range 0.15<pT<10 GeV/c. The correlation between transverse momentum and particle multiplicity is also studied. Results are presented for inelastic (INEL) and non-single-diffractive (NSD) events. The average transverse momentum for |η|<0.8 is 〈pT〉INEL=0.483±0.001 (stat.)±0.007 (syst.) GeV/c and 〈pT〉NSD=0.489±0.001 (stat.)±0.007 (syst.) GeV/c, respectively. The data exhibit a slightly larger 〈pT〉 than measurements in wider pseudorapidity intervals. The results are compared to simulations with the Monte Carlo event generators PYTHIA and PHOJET.
Harmonic decomposition of two particle angular correlations in Pb–Pb collisions at √sNN=2.76 TeV

Angular correlations between unidentified charged trigger (t) and associated (a) particles are measured by the ALICE experiment in Pb–Pb collisions at √sNN=2.76 TeV for transverse momenta 0.25<pTt,a<15 GeV/c, where pTt>pTa. The shapes of the pair correlation distributions are studied in a variety of collision centrality classes between 0 and 50% of the total hadronic cross section for particles in the pseudorapidity interval |η|<1.0. Distributions in relative azimuth Δϕ≡ϕt−ϕa are analyzed for |Δη|≡|ηt−ηa|>0.8, and are referred to as “long-range correlations”. Fourier components VnΔ≡〈cos(nΔϕ)〉 are extracted from the long-range azimuthal correlation functions. If particle pairs are correlated to one another through their individual correlation to a common symmetry plane, then the pair anisotropy VnΔ(pTt,pTa) is fully described in terms of single-particle anisotropies vn(pT) as VnΔ(pTt,pTa)=vn(pTt)vn(pTa). This expectation is tested for 1⩽n⩽5 by applying a global fit of all VnΔ(pTt,pTa) to obtain the best values vn{GF}(pT). It is found that for 2⩽n⩽5, the fit agrees well with data up to pTa∼3–4 GeV/c, with a trend of increasing deviation as pTt and pTa are increased or as collisions become more peripheral. This suggests that no pair correlation harmonic can be described over the full 0.25<pT<15 GeV/c range using a single vn(pT) curve; such a description is however approximately possible for 2⩽n⩽5 when pTa<4 GeV/c. For the n=1 harmonic, however, a single v1(pT) curve is not obtained even within the reduced range pTa<4 GeV/c.
Rapidity and transverse momentum dependence of inclusive J/ψ production in pp collisions at √s=7 TeV

The ALICE experiment at the LHC has studied inclusive J/ψ production at central and forward rapidities in pp collisions at √s=7 TeV. In this Letter, we report on the first results obtained detecting the J/ψ through the dilepton decay into e+e− and μ+μ− pairs in the rapidity ranges |y|<0.9 and 2.5<y<4, respectively, and with acceptance down to zero pT. In the dielectron channel the analysis was carried out on a data sample corresponding to an integrated luminosity Lint=5.6 nb−1 and the number of signal events is NJ/ψ=352±32(stat.)±28(syst.); the corresponding figures in the dimuon channel are Lint=15.6 nb−1 and NJ/ψ=1924±77(stat.)±144(syst.). The measured production cross sections are σJ/ψ(|y|<0.9)=10.7±1.0(stat.)±1.6(syst.)−2.3+1.6(syst.pol.)μb and σJ/ψ(2.5<y<4)=6.31±0.25(stat.)±0.76(syst.)−1.96+0.95(syst.pol.)μb. The differential cross sections, in transverse momentum and rapidity, of the J/ψ were also measured.
The first measurement of two-pion Bose–Einstein correlations in central Pb–Pb collisions at √sNN=2.76 TeV at the Large Hadron Collider is presented. We observe a growing trend with energy now not only for the longitudinal and the outward but also for the sideward pion source radius. The pion homogeneity volume and the decoupling time are significantly larger than those measured at RHIC.
Inclusive transverse momentum spectra of primary charged particles in Pb–Pb collisions at √sNN=2.76 TeV have been measured by the ALICE Collaboration at the LHC. The data are presented for central and peripheral collisions, corresponding to 0–5% and 70–80% of the hadronic Pb–Pb cross section. The measured charged particle spectra in |η|<0.8 and 0.3<pT<20 GeV/c are compared to the expectation in pp collisions at the same sNN, scaled by the number of underlying nucleon–nucleon collisions. The comparison is expressed in terms of the nuclear modification factor RAA. The result indicates only weak medium effects (RAA≈0.7) in peripheral collisions. In central collisions, RAA reaches a minimum of about 0.14 at pT=6–7 GeV/c and increases significantly at larger pT. The measured suppression of high-pT particles is stronger than that observed at lower collision energies, indicating that a very dense medium is formed in central Pb–Pb collisions at the LHC.
Das humane Immundefizienz-Virus (HIV) benötigt für die Virus-Zellbindung spezifische Oberflächenrezeptoren auf den Wirtszellen (z. B. CD4, CXCR4, CCR5). Zurzeit basiert die Behandlung der chronisch persistierenden HIV-Erkrankung auf einer lebenslangen Chemotherapie (Highly Active Antiretroviral Therapy, ART) bestehend beispielsweise aus einer Kombination von 2 Nukleosidanaloga und einem Protease-Inhibitor, die das Virus nicht eradiziert, sondern nur in seiner Vermehrung hemmt. Dies birgt jedoch die Gefahr der Entwicklung von Resistenzen gegenüber der medikamentösen Therapie. Zusätzlich wird eine Veränderung der HIV-Rezeptorspezifität unter der Behandlung mit Antagonisten des HIV-Rezeptors CCR5 befürchtet. Cytarabin (Ara-C) ist ein Zytostatikum, das in der Therapie von Leukämien eingesetzt wird. Als Nukleosidanalogon gehört es strukturell zur selben Wirkstoffklasse wie die in der HIV-Therapie eingesetzten Nukleosidanaloga, jedoch sind bisher keine antiretroviralen Eigenschaften für Ara-C beschrieben worden. Die T-lymphoide Zelllinie C8166 ist permissiv für HIV. Die Adaptation von C8166-Zellen an das Wachstum in Gegenwart von Ara-C (Zellinie C8166rAra-C5μM) resultierte in einer signifikanten Verringerung der Oberflächenexpression der HIV-Rezeptoren CD4 und CXCR4 und zu einer verringerten Permissivität gegenüber HIV. In der vorliegenden Arbeit sollte untersucht werden, ob die Adaptation an Ara-C bei anderen T-lymphoiden Zelllinien ebenfalls zur Verringerung der Expression von CD4, und CXCR4 führt. Zusätzlich sollte untersucht werden, wie sich die Expression von CCR5 verhält. Es wurden die folgenden parentalen und an Ara-C adaptieten T-lymphoiden Zelllinien verwendet: H9, H9rAra-C600μM, MOLT4/8, MOLT4/8rAra-C100μM und MOLT4/8rAra-C200μM. Bei allen Ara-C resistenten Zelllinien kam es zu einer signifikant verringerten Expression von CD4 und CXCR4 auf mRNA und Proteinebene sowie zu einer signifikanten Erhöhung der CCR5-Expression. Im Gegensatz hierzu zeigten an AZT adaptierte H9-Zellen (H9rAZT3000μM) keine signifikante Veränderung in der Expression von CD4, CXCR4 oder CCR5 im Vergleich zu parentalen H9-Zellen. Die akute Behandlung der parentalen H9-Zellen mit niedrigen, untoxischen Ara-C Konzentrationen führte ebenfalls zu einem Anstieg der CCR5-Expression und zu einer Verminderung der CD4- und CXCR4-Expression. Zellzyklusmessungen ergaben, dass der Zellzyklus in mit untoxischen Ara-C-Konzentrationen behandelten H9-Zellen (Anstieg der Zellteilungsrate auf das 2-fache) und in allen an Ara-C adaptierten Zelllinien im Vergleich zu den unbehandelten bzw. parentalen Zellen stärker stimuliert war. Epigenetische Einflüsse könnten bei der veränderten Expression von CD4, CXCR4 und/oder CCR5 in Ara-C resistenten Zellen eine Rolle spielen. Dies erscheint jedoch unwahrscheinlich, da weder der DNA-Methylierungsinhibitor Aza-C noch der Histondeacetylase-Inhibitor SAHA die Expression von CD4, CXCR4 oder CCR5 beeinflussten. Weitere Untersuchungen müssen zeigen, ob eine Kombination von Ara-C, das zu einer Verringerung der CXCR4- und CD4-Expression und zu einer Erhöhung der CCR5-Expression führt, mit CCR5-Inhibitoren eine therapeutische Option darstellt. Möglicherweise wirkt die Verwendung von Ara-C auch einem CCR5/CXCR4-Shift entgegen.
Phase transitions in a non-perturbative regime can be studied by ab initio Lattice Field Theory methods. The status and future research directions for LFT investigations of Quantum Chromo-Dynamics under extreme conditions are reviewed, including properties of hadrons and of the hypothesized QCD axion as inferred from QCD topology in different phases. We discuss phase transitions in strong interactions in an extended parameter space, and the possibility of model building for Dark Matter and Electro-Weak Symmetry Breaking. Methodological challenges are addressed as well, including new developments in Artificial Intelligence geared towards the identification of different phases and transitions.
Die vorliegende Dissertationsschrift befasst sich mit der molekulargenetischen Analyse zweier Basalganglienerkrankungen. Zum einen wurden Patienten mit M. Parkinson genetisch untersucht, zum anderen Patienten mit autosomal dominanter zervikaler Torsionsdystonie. Die Aufgabe bestand in der passenden Wahl der Methode zur jeweiligen humangenetischen Fragestellung. Der erste Teil handelte von der Suche der krankheitsverursachenden Mutation für die autosomal dominante zervikale Torsionsdystonie mit Spätmanifestation auf Chromosom 18p (Kandidatenlokus DYT7). Die erkrankte Familie deutscher Herkunft zeigt dystone Symptome mit Betonung auf kraniozervikale und brachiale Körperabschnitte und ist somit die weltweit einzige bekannte Familie mit Vererbung dieser ansonsten sporadisch auftretenden Erkrankung. Die PCR-Sequenzierung der Kandidatengene ZFP161, LOC390828, NDUFV2 und PTPRM auf dem DYT7 Lokus erbrachte bei den sieben erkrankten Familienmitgliedern im Vergleich zu nicht verwandten Kontrollen (Ehepartner und 96 Kontrollen der Blutbank) keinen Aminosäureaustausch, der ausschließlich bei den erkrankten Probanden zu finden war. Technisch konzentrierte sich diese Untersuchung auf die Amplifizierung und anschließende Sequenzierung jedes einzelnen Exons in den zu untersuchenden Proben, und die Bestätigung einer putativen Mutation mittels Verdau der PCR-Produkte durch Restriktionsendonukleasen. Die Auswahl der Kandidatengene erfolgte aufgrund der Annahme pathobiochemischer Mechanismen, die durch andere Formen der vererbten Torsionsdystonie oder zellbiologische Experimente als krankheitsverursachend gelten. Auch wenn keine Mutation gefunden wurde, so konnten bereits bekannte und neue single nucleotide polymorphisms (SNP) etabliert werden. Die zweite Thematik befasste sich mit der Frage, ob das bereits bekannte Parkinson-Gen UCH-L1 auf dem PARK5 Lokus krankheitsverursachend für den autosomal dominanten M. Parkinson in einer spanischen Familie ist. Diese parametrische Kopplungsanalyse wurde mithilfe der heißen Polyacrylamid-Gelelektrophorese (PAGE) durchgeführt. Dabei konnte in allen Patienten und den Verwandten ersten Grades über Analyse der Mikrosatelliten nördlich und südlich der Kandidatenregion (UCH-L1) in einem Bereich sehr niedriger Rekombinationswahrscheinlichkeit eine Haplotypisierung erfolgen. Die Haplotypisierung zeigte nicht die erforderliche Identifizierung eines Krankheitsallels bei allen betroffenen Probanden. Somit ist hier neben der einzig bekannten deutschen PARK5 Familie keine weitere Familie mit UCH-L1 Mutation bestätigt worden. Dementsprechend ist die Ätiologie dieser Erkrankung in dieser Familie noch unklar, was aber der Bedeutung des Ubiquitin-Proteasom Systems in der Parkinson-Entität keinen Abbruch getan hat. Da alle anderen autosomal dominanten Parkinson-Loci ausgeschlossen sind, muss sich die Ursache für den M. Parkinson in dieser Familie in einem heute noch unbekannten Gen befinden. Weitere Untersuchungen im Rahmen eines Genomscans sind aufgrund der geringen Fallzahl nicht möglich. Die letzte Aufgabe dieser Arbeit bestand in der Durchführung einer Assoziationsstudie mit den putativen PINK1 (PARK6) Interaktoren NME4 und MTIF3 für den mehrheitlich sporadisch auftretenden M. Parkinson. Dabei wurden in zwei unabhängigen Studiengruppen mit insgesamt 453 sporadischen Parkinsonpatienten und 370 Kontrollen jeweils zwei SNPs auf gekoppelte Vererbung mit der Erkrankung untersucht. Der Unterschied zwischen den Testgruppen bestand im Studiendesign, da zum einen mit den Patienten nicht verwandte Kontrollen und zum anderen verwandte Kontrollen verwendet wurden. Die mit beiden Studientypen normalerweise auftretenden Probleme durch Stratifikation bzw. erniedrigte statistische Power konnten durch Kombination der Studien ausgeglichen werden. Das Methodenspektrum umfasste PCR und Restriktionsverdau, was zum Auffinden eines Kopplungsungleichgewichts für das Gen MTIF3 führte. Ein heterozygoter Basenaustausch für den Polymorphismus rs7669 erhöht signifikant das Relative Risiko an M. Parkinson zu erkranken, wohingegen der homozygote Basenaustausch das Krankheitsrisiko des Trägers signifikant erniedrigt. Bezüglich des Relativen Risikos wurde der Effekt der molekularen Heterosis nachgewiesen. Bei diesem mitochondrial lokalisierten Gen handelt es sich um einen Initiator der mitochondrialen Translation. Demzufolge besteht hier Einfluss auf die Homöostase und somit Funktionalität der Atmungskettenkomplexe, die als bedeutend für die Pathogenese des M. Parkinson angesehen werden. Die Verbindung zum mitochondrial lokalisierten PINK1 besteht aufgrund seiner Kinase-Aktivität in der An- und Abschaltung des mitochondrialen Translations - Initiationsfaktors. Aber auch die Wichtigkeit von NME4 konnte in dieser Studie trotz fehlender Assoziation nicht ausgeschlossen werden, da vorangehende experimentelle Ergebnisse dieses Protein bereits in den PINK1 Signalweg zuordnen konnten. MTIF3 könnte wohlmöglich ein wichtiger genetische Risikofaktor für den idiopathischen M. Parkinson sein. Es bleibt abzuwarten, ob zukünftige genetische und zellbiologische Experimente die Wichtigkeit, die in diesem Protein zu liegen scheint, bestätigen können.
We show the existence of additive kinematic formulas for general flag area measures, which generalizes a recent result by Wannerer. Building on previous work by the second named author, we introduce an algebraic framework to compute these formulas explicitly. This is carried out in detail in the case of the incomplete flag manifold consisting of all (p+1)-planes containing a unit vector.
We show how nonlocal boundary conditions of Robin type can be encoded in the pointwise expression of the fractional operator. Notably, the fractional Laplacian of functions satisfying homogeneous nonlocal Neumann conditions can be expressed as a regional operator with a kernel having logarithmic behaviour at the boundary.
We provide a Hopf boundary lemma for the regional fractional Laplacian (−Δ)sΩ, with Ω ⊂ RN a bounded open set. More precisely, given u a pointwise or weak super-solution of the equation (−Δ)s u = c(x)u in Ω, we show that the ratio u(x)∕(dist(x, 𝜕Ω))2s−1 is strictly Ω positive as x approaches the boundary 𝜕Ω of Ω. We also prove a strong maximum principle for distributional super-solutions.
We show explicit formulas for the evaluation of (possibly higher-order) fractional Laplacians (-△)ˢ of some functions supported on ellipsoids. In particular, we derive the explicit expression of the torsion function and give examples of s-harmonic functions. As an application, we infer that the weak maximum principle fails in eccentric ellipsoids for s ∈ (1; √3 + 3/2) in any dimension n ≥ 2. We build a counterexample in terms of the torsion function times a polynomial of degree 2. Using point inversion transformations, it follows that a variety of bounded and unbounded domains do not satisfy positivity preserving properties either and we give some examples.

Clean water is fundamental to human health and ecosystem integrity. However, water quality deteriorates due to novel anthropogenic pollutants present at microgram per liter concentrations in urban water cycles (termed micropollutants). Wastewater treatment plants (WWTP) have been identified as major point sources for aquatic (micro-)pollutants. Chemical and ecotoxicological analyses have shown that conventional biological WWTPs do not fully remove micropollutants and associated toxicities, which is often because of mobile, polar and/or recalcitrant compounds and transformation products (TPs). To minimize possible environmental risks, advanced wastewater treatment (AWWT) technologies could be a promising mitigation measure. Multiple processes are therefore being developed and evaluated such as ozonation and ozonation followed by granulated activated carbon (GAC) or biological filtration. Assessing the performance of these combined AWWTs was the focus the TransRisk project. Within this project, this thesis accomplished four major goals.
Firstly, the preparation of (waste)water samples was optimised for in vitro bioassays. Acidification, filtration and solid phase extraction (SPE) were tested for their impact on environmentally relevant in vitro endocrine activities, mutagenicity, genotoxicity and cytotoxicity. Significantly different outcomes of these assays were detected comparing neutral and acidified samples. Sample filtration had a lesser impact, but in some cases retention of particle-bound compounds could have caused significant toxicity losses. Out of three SPE sorbents the Telos C18/ENV at sample pH 2.5 extracted highest toxicity, some undetected in aqueous samples. These results indicate that sample preparation needs to be optimised for specific sample matrices and bioassays to avoid false-positive or -negative detects in effect-based analyses.
Secondly, the above listed in vitro toxicities were monitored in a protected region for drinking water production in South-West Germany (2012-2015). Out of 30 sampling sites surface water and groundwater were the least polluted. Nonetheless, a few groundwater samples induced high anti-estrogenic activity that prompted further monitoring. The latter included a waterworks in which no toxicity was detected. Hospital wastewater also had elevated in vitro toxicities and hospitals are, thus, relevant intervention points for source control. The biological WWTPs were effective in removing most of the detected toxicity, and the selected bioassays proved to be pertinent tools for water quality assessment and prioritisation of pollution hotspots.
Thirdly, the in vivo bioassay ISO10872 based on Caenorhabditis elegans (C. elegans) was adapted for this thesis. Using this model, a median effect concentration (EC50) for reproductive toxicity of the polycyclic aromatic hydrocarbon β-naphthoflavone (β- NF) of 114 µg/L was computed which is slightly lower than reported in the scientific literature. β-NF induced cyp-35A3::GFP (a biomarker in transgenic animals) in a time and concentration dependent manner (≤ 21.3–24 fold above controls). β-NF spiked wastewater samples supported earlier hypotheses on particle-bound pollutants. Reproductive toxicity (96 h) and cyp-35A3 induction (24 h) of biologically treated and/or ozonated wastewater extracts and growth promoting effects of GAC/biologically filtered ozonated wastewater extracts were observed. This suggested the presence of residual bioactive/toxic chemicals not included in the targeted chemical analysis. It also highlighted the importance of integrating multiple (apical and molecular) endpoints in wastewater assessments.
Fourthly, five in vitro and the adapted C. elegans bioassay were integrated into a wastewater quality evaluation (developed within TransRisk). Out of the five AWWT options, ozonation (at 1 g O3,applied/g DOC, HRT ~ 18 min) combined with nonaerated GAC filtration was rated most effective for toxicity removal. All five AWWTs largely removed estrogenic and (anti-)androgenic activities, but not anti-estrogenic activity and mutagenicity, which even increased during ozonation. This has been observed in related studies and points towards toxic TPs. These results also emphasized the need for implementing an effective post-treatment for ozonation. The results from a parallel in vivo study with Lumbriculus variegatus and Potamopyrgus antipodarum conducted on site at the WWTP (using flow through systems) were in accordance with the C. elegans results. In this context, it is suggested to further implement C. elegans as sensitive, feasible and ecologically relevant model.
In conclusion, this thesis shows how optimised sample preparation, long-term (in vitro) environmental monitoring, sensitive and ecologically relevant (in vivo) bioassays as well as innovative evaluation concepts, are pivotal in improving the removal of micropollutants and their toxicities with AWWTs. Future research should further develop and evaluate measures at sewer systems, conventional biological, tertiary and other advanced treatment technologies, as well as sociopolitical strategies (e.g., source control or natural conservation) and restoration projects. The effect-based tools optimised in this thesis will support assessing their success.

Das Hypophysenadenom als Ursache einer sekundären Nebennierenrindeninsuffizienz, nur mit isoliertem ACTH-Defizit, ist außergewöhnlich. Ein ACTH-Mangel tritt in der Regel nicht isoliert, sondern zusammen mit dem Ausfall anderer Hypophysenfunktionen auf. Besonders bei Patienten mit Kinderwunsch sollte den Erkrankungen der Hypophyse und der Nebennieren große Aufmerksamkeit geschenkt werden.
Bacterial biosynthetic assembly lines, such as non-ribosomal peptide synthetases (NRPS) and polyketide synthases, are often subject of synthetic biology – because they produce a variety of natural products invaluable for modern pharmacotherapy. Acquiring the ability to engineer these biosynthetic assembly lines allows the production of artificial non-ribosomal peptides (NRP), polyketides, and hybrids thereof with new or improved properties. However, traditional bioengineering approaches have suffered for decades from their very limited applicability and, unlike combinatorial chemistry, are stigmatized as inefficient because they cannot be linked to the high-throughput screening platforms of the pharmaceutical industry. Although combinatorial chemistry can generate new molecules cheaper, faster, and in greater numbers than traditional natural product discovery and bioengineering approaches, it does not meet current medical needs because it covers only a limited biologically relevant chemical space. Hence, methods for high-throughput generation of new natural product-like compound libraries could provide a new avenue towards the identification of new lead compounds. To this end, prior to this work, we introduced an artificial synthetic NRPS type, referred to as type S NRPS, to provide a first-of-its-kind bicombinatorial approach to parallelized high-throughput NRP library generation. However, a bottleneck of these first two generations of type S NRPS was a significant drop in production yields. To address this issue, we applied an iterative optimization process that enabled titer increases of up to 55-fold compared to the non-optimized equivalents, restoring them to wild-type levels and beyond.

In high-energy heavy-ion collisions, partonic collectivity is evidenced by the constituent quark number scaling of elliptic flow anisotropy for identified hadrons. A breaking of this scaling and dominance of baryonic interactions is found for identified hadron collective flow measurements in √sNN = 3 GeV Au+Au collisions. In this paper, we report measurements of the first- and second-order azimuthal anisotropic parameters, v1 and v2, of light nuclei (d, t, 3He, 4He) produced in √sNN = 3 GeV Au+Au collisions at the STAR experiment. An atomic mass number scaling is found in the measured v1 slopes of light nuclei at mid-rapidity. For the measured v2 magnitude, a strong rapidity dependence is observed. Unlike v2 at higher collision energies, the v2 values at mid-rapidity for all light nuclei are negative and no scaling is observed with the atomic mass number. Calculations by the Jet AA Microscopic Transport Model (JAM), with baryonic mean-field plus nucleon coalescence, are in good agreement with our observations, implying baryonic interactions dominate the collective dynamics in 3 GeV Au+Au collisions at RHIC.
We report the first multi-differential measurements of strange hadrons of K −, φ and − yields as well as the ratios of φ/K − and φ/− in Au+Au collisions at √sNN = 3 GeV with the STAR experiment fixed target configuration at RHIC. The φ mesons and − hyperons are measured through hadronic decay channels, φ → K + K − and Ξ− → Λπ−. Collision centrality and rapidity dependence of the transverse momentum spectra for these strange hadrons are presented. The 4π yields and ratios are compared to thermal model and hadronic transport model predictions. At this collision energy, thermal model with grand canonical ensemble (GCE) under-predicts the φ/K − and φ/− ratios while the result of canonical ensemble (CE) calculations reproduce φ/K −, with the correlation length rc ∼ 2.7 fm, and φ/−, rc ∼ 4.2 fm, for the 0-10% central collisions. Hadronic transport models including high mass resonance decays could also describe the ratios. While thermal calculations with GCE work well for strangeness production in high energy collisions, the change to CE at 3 GeV implies a rather different medium property at high baryon density.
We report on the measurements of directed flow v1 and elliptic flow v2 for hadrons (π±, K ±, K0 S , p, φ, Λ and ) from Au+Au collisions at √sN N = 3 GeV and v2 for (π±, K ±, p and p) at 27 and 54.4 GeV with the STAR experiment. While at the two higher energy midcentral collisions the numberof-constituent-quark (NCQ) scaling holds, at 3 GeV the v2 at midrapidity is negative for all hadrons and the NCQ scaling is absent. In addition, the v1 slopes at midrapidity for almost all observed hadrons are found to be positive, implying dominant repulsive baryonic interactions. The features of negative v2 and positive v1 slope at 3 GeV can be reproduced with a baryonic mean-field in transport model calculations. These results imply that the medium in such collisions is likely characterized by baryonic interactions.
Measurement of cold nuclear matter effects for inclusive J/ψ in p+Au collisions at √sNN = 200 GeV

Measurement by the STAR experiment at RHIC of the cold nuclear matter (CNM) effects experienced by inclusive J/ψ at mid-rapidity in 0-100% p+Au collisions at √sNN = 200 GeV is presented. Such effects are quantified utilizing the nuclear modification factor, RpAu, obtained by taking a ratio of J/ψ yield in p+Au collisions to that in p+p collisions scaled by the number of binary nucleon-nucleon collisions. The differential J/ψ yield in both p+p and p+Au collisions is measured through the dimuon decay channel, taking advantage of the trigger capability provided by the Muon Telescope Detector in the RHIC 2015 run. Consequently, the J/ψ RpAu is derived within the transverse momentum (pT) range of 0 to 10 GeV/c. A suppression of approximately 30% is observed for pT < 2 GeV/c, while J/ψ RpAu becomes compatible with unity for pT greater than 3 GeV/c, indicating the J/ψ yield is minimally affected by the CNM effects at high pT. Comparison to a similar measurement from 0-20% central Au+Au collisions reveals that the observed strong J/ψ suppression above 3 GeV/c is mostly due to the hot medium effects, providing strong evidence for the formation of the quark-gluon plasma in these collisions. Several model calculations show qualitative agreement with the measured J/ψ RpAu, while their agreement with the J/ψ yields in p+p and p+Au collisions is worse.
Azimuthal anisotropy measurement of (multi-)strange hadrons in Au+Au collisions at √sNN = 54.4 GeV

Azimuthal anisotropy of produced particles is one of the most important observables used to access the collective properties of the expanding medium created in relativistic heavy-ion collisions. In this paper, we present second (v2) and third (v3) order azimuthal anisotropies of K0S, ϕ, Λ, Ξ and Ω at mid-rapidity (|y|<1) in Au+Au collisions at sNN−−−√ = 54.4 GeV measured by the STAR detector. The v2 and v3 are measured as a function of transverse momentum and centrality. Their energy dependence is also studied. v3 is found to be more sensitive to the change in the center-of-mass energy than v2. Scaling by constituent quark number is found to hold for v2 within 10%. This observation could be evidence for the development of partonic collectivity in 54.4 GeV Au+Au collisions. Differences in v2 and v3 between baryons and anti-baryons are presented, and ratios of v3/v3/22 are studied and motivated by hydrodynamical calculations. The ratio of v2 of ϕ mesons to that of anti-protons (v2(ϕ)/v2(p¯)) shows centrality dependence at low transverse momentum, presumably resulting from the larger effects from hadronic interactions on anti-proton v2.
We report the measurement of K∗0 meson at midrapidity (|y|< 1.0) in Au+Au collisions at sNN−−−√~=~7.7, 11.5, 14.5, 19.6, 27 and 39 GeV collected by the STAR experiment during the RHIC beam energy scan (BES) program. The transverse momentum spectra, yield, and average transverse momentum of K∗0 are presented as functions of collision centrality and beam energy. The K∗0/K yield ratios are presented for different collision centrality intervals and beam energies. The K∗0/K ratio in heavy-ion collisions are observed to be smaller than that in small system collisions (e+e and p+p). The K∗0/K ratio follows a similar centrality dependence to that observed in previous RHIC and LHC measurements. The data favor the scenario of the dominance of hadronic re-scattering over regeneration for K∗0 production in the hadronic phase of the medium.

We report high-precision measurements of the longitudinal double-spin asymmetry, 𝐴𝐿𝐿, for midrapidity inclusive jet and dijet production in polarized 𝑝𝑝 collisions at a center-of-mass energy of √𝑠=200 GeV. The new inclusive jet data are sensitive to the gluon helicity distribution, Δ𝑔(𝑥,𝑄2), for gluon momentum fractions in the range from 𝑥≃0.05 to 𝑥≃0.5, while the new dijet data provide further constraints on the 𝑥 dependence of Δ𝑔(𝑥,𝑄2). The results are in good agreement with previous measurements at √𝑠=200 GeV and with recent theoretical evaluations of prior world data. Our new results have better precision and thus strengthen the evidence that Δ𝑔(𝑥,𝑄2) is positive for 𝑥>0.05.
Rationale: The AMP-activated protein kinase (AMPK) is stimulated by hypoxia, and although the AMPKα1 catalytic subunit has been implicated in angiogenesis, little is known about the role played by the AMPKα2 subunit in vascular repair.
Objective: To determine the role of the AMPKα2 subunit in vascular repair.
Methods and Results: Recovery of blood flow after femoral artery ligation was impaired (>80%) in AMPKα2-/- versus wild-type mice, a phenotype reproduced in mice lacking AMPKα2 in myeloid cells (AMPKα2ΔMC). Three days after ligation, neutrophil infiltration into ischemic limbs of AMPKα2ΔMC mice was lower than that in wild-type mice despite being higher after 24 hours. Neutrophil survival in ischemic tissue is required to attract monocytes that contribute to the angiogenic response. Indeed, apoptosis was increased in hypoxic neutrophils from AMPKα2ΔMC mice, fewer monocytes were recruited, and gene array analysis revealed attenuated expression of proangiogenic proteins in ischemic AMPKα2ΔMC hindlimbs. Many angiogenic growth factors are regulated by hypoxia-inducible factor, and hypoxia-inducible factor-1α induction was attenuated in AMPKα2-deficient cells and accompanied by its enhanced hydroxylation. Also, fewer proteins were regulated by hypoxia in neutrophils from AMPKα2ΔMC mice. Mechanistically, isocitrate dehydrogenase expression and the production of α-ketoglutarate, which negatively regulate hypoxia-inducible factor-1α stability, were attenuated in neutrophils from wild-type mice but remained elevated in cells from AMPKα2ΔMC mice.
Conclusions: AMPKα2 regulates α-ketoglutarate generation, hypoxia-inducible factor-1α stability, and neutrophil survival, which in turn determine further myeloid cell recruitment and repair potential. The activation of AMPKα2 in neutrophils is a decisive event in the initiation of vascular repair after ischemia.
Medicinal plants represent a big reservoir for discovering new drugs against all kinds of diseases including inflammation. In spite the large number of promising anti-inflammatory plant extracts and isolated components, research on medicinal plants proves to be very difficult. Based on that background this review aims to provide a summarized insight into the hitherto known pharmacologically active concentrations, bioavailability, and clinical efficacy of boswellic acids, curcumin, quercetin and resveratrol. These examples have in common that the achieved plasma concentrations were found to be often far below the determined IC50 values in vitro. On the other hand demonstrated therapeutic effects suggest a necessity of rethinking our pharmacokinetic understanding. In this light this review discusses the value of plasma levels as pharmacokinetic surrogates in comparison to the more informative value of tissue concentrations. Furthermore the need for new methodological approaches is addressed like the application of combinatorial approaches for identifying and pharmacokinetic investigations of active multi-components. Also the physiological relevance of exemplary in vitro assays and absorption studies in cell-line based models is discussed. All these topics should be ideally considered to avoid inaccurate predictions for the efficacy of herbal components in vivo and to unlock the “black box” of herbal mixtures.
Dosing accuracy of two disposable insulin pens according to new ISO 11608-1: 2012 requirements.

OBJECTIVE: The aim was to compare 2 disposable insulin pens, FlexTouch® (Novo Nordisk, insulin aspart) and SoloSTAR® (Sanofi, insulin glulisine), according to new ISO 11608-1:2012 requirements for dosing accuracy.
METHODS: Sixty pens of each type were tested at 1, 40, and 80 U doses. Following the new ISO requirements, each dose was delivered from the front, middle, and rear one-third of the pen. Statistical analysis was performed using Student's t test.
RESULTS: Both pens delivered all doses within ISO limits. The difference between the average measured dose and the target dose was significantly smaller for SoloSTAR than FlexTouch at 40 U (P = .009) and 80 U (P = .008), but not at 1 U (P = .417).
CONCLUSION: Both insulin pens fulfilled the dosing accuracy requirements defined by ISO 11608-1:2012 at all 3 dosage levels.
Optimal investment decisions by institutional investors require accurate predictions with respect to the development of stock markets. Motivated by previous research that revealed the unsatisfactory performance of existing stock market prediction models, this study proposes a novel prediction approach. Our proposed system combines Artificial Intelligence (AI) with data from Virtual Investment Communities (VICs) and leverages VICs’ ability to support the process of predicting stock markets. An empirical study with two different models using real data shows the potential of the AI-based system with VICs information as an instrument for stock market predictions. VICs can be a valuable addition but our results indicate that this type of data is only helpful in certain market phases.
Artificial Intelligence (AI) and Machine Learning (ML) are currently hot topics in industry and business practice, while management-oriented research disciplines seem reluctant to adopt these sophisticated data analytics methods as research instruments. Even the Information Systems (IS) discipline with its close connections to Computer Science seems to be conservative when conducting empirical research endeavors. To assess the magnitude of the problem and to understand its causes, we conducted a bibliographic review on publications in high-level IS journals. We reviewed 1,838 articles that matched corresponding keyword-queries in journals from the AIS senior scholar basket, Electronic Markets and Decision Support Systems (Ranked B). In addition, we conducted a survey among IS researchers (N = 110). Based on the findings from our sample we evaluate different potential causes that could explain why ML methods are rather underrepresented in top-tier journals and discuss how the IS discipline could successfully incorporate ML methods in research undertakings.
Objective: The aim of this study was to report the basic cerebrospinal fluid (CSF) profile in patients with primary progressive multiple sclerosis (PPMS).
Methods: The results of CSF analysis from 254 patients with PPMS were collected at four university hospitals in Germany. Routine CSF parameters and different indices of intrathecal immunoglobulin synthesis were evaluated. We assessed possible correlations between the various CSF parameters and the expanded disability status scale (EDSS) both at the time of lumbar puncture and during the course of the disease.
Results: The median cell count and albumin concentration in the CSF did not deviate from normal values. The CSF-serum albumin-quotient (QALB) was elevated in 29.6% of the patients, while intrathecal immunoglobulin G (IgG) oligoclonal bands (OCBs) were detected in 91.1% of the patients. CSF-lactate levels as well as local IgM- and IgA-synthesis were correlated with the yearly disease progression rate, as assessed by EDSS.
Conclusion: We present the results of the hitherto largest and most detailed CSF biomarker profile in a cohort of 254 patients with PPMS. As reported previously, OCBs are the most sensitive marker for intrathecal IgG synthesis. CSF-lactate concentrations are positively correlated with the progression rate, which might suggest that mitochondrial dysfunction plays a relevant role in PPMS. The negative correlation between intrathecally produced IgM and IgA and disease progression may indicate their hitherto unexplored protective role.
18-OH-Corticosteron (18 ) wird als die unmittelbare Vorstufe der Aldosteron-Synthese angesehen. In-vitro-Untersuchungen sowie vereinzelten klinischen Beobachtungen zufolge sollen Nebennierenrinden-Adenome, im Gegensatz zu Nebennierenrinden-Hyperplasie, vermehrt 8- bilden. In der vorliegenden Studie wurde an 1.272 Patienten einer Hochdruckambulanz, wobei bei 84 Patienten mit primärem Aldosteronismus infolge eines Adenoms sowie bei 110 Patienten infolge einer Nebennierenrinden-Hyperplasie die Diagnose gesichert werden konnte, der diagnostische Stellenwert von 8- im Vergleich zu den Aldosteron-Metaboliten Aldosteron-18-Glucuronid (ALD-18-G) und Tetrahydroaldosteron (TH-ALD) untersucht. Dies im Hin blick auf: 1. die Erkennung eines primären Aldosteronismus, und 2. der differentialdiagnostischen Unterscheidung zwischen einem Adenom und einer Hyperpläsie.
Bezüglich der ersten Fragestellung wurde für 18-OHB - hinsichtlich der Unterscheidung zwischen dem primären Aldosteronismus infolge eines Adenoms und einer essentiellen Hypertonie-eine diagnostische Sensitivität von 99,2% bei einer diagnostischen Spezifität von 95,2% berechnet. Deutlich geringer war mit einer diagnostischen Sensitivität von 79,7% bei einer diagnostischen Spezifität von 60,9% die Abgrenzung zwischen dem primären Aldosteronismus infolge einer Hyperpläsie und einer essentiellen Hypertonie.
18-OHB war bei 11 der 84 Adenom- und 5 der 110 Hyperplasie-Patienten zunächst das einzig erhöhte Steroid im 24 h-Urin. Bei ihnen konnte erst innerhalb einer bis zu 2jährigen Beobachtungszeit ein langsamer Anstieg der Aldosteron-Metabolite beobachtet werden. Somit stellt das 18-OHB einen „Frühmarker" der Erkrankung dar.
In der Unterscheidung zwischen einem Nebennierenrinden-Adenom und einer -Hyperplasie besitzt 18-OHB mit einer diagnostischen Sensitivität von 84,5% bei einer diagnostischen Spezifität von 96,4% ein höheres Abgrenzungsvermögen als Tetrahydro-Aldosteron und Aldosteron-18-Glucuronid dar.
Für die Unterscheidung des Adenoms von der Hyperpläsie ließ sich für 18-OH-Corticosteron im 24 h-Urin ein Wert von 7,9 [ig/die als eine optimale Diskriminanzschwelle berechnen. Bei einer höheren Ausscheidung ist in 84,5% der Fälle mit einem Nebennierenrinden-Adenom zu rechnen. Hinsichtlich der diagnostischen Wertung von 18-OHB sind weder geschlechts- noch altersspezifische Abhängigkeiten zu berücksichtigen.
Die Bestimmung von Tetrahydro-Aldosteron (oder Aldosteron-18-Glucuronid) in Kombination mit 18-OH-Corticosteron stellt die optimale Methode zur Diagnostik des primären Hyperaldosteronismus, insbesondere infolge eines Adenoms, dar.
The Crusade movement is one of the most important occurrences of medieval history. It took place throughout two centuries in the Levant and affected both Muslims and Crusaders and in turn changed the way in which West and East related to one another.1 When the Crusaders took control of the Holy Land and many Islamic cities in the Levant, they transferred their feudal European system there. They established four main fiefdoms or lordships, Jerusalem, Edessa, Antioch and Tripoli. In addition, there were another twelve secondary fiefdoms,2 of which Tibnīn was one. Tibnīn was called “Toron” by the Crusaders. Once the Crusaders had captured Tibnīn, they began building its fortified castle, from which the fief of Tibnīn gained its importance throughout the period of the Crusades.
This paper traces the military role of Tibnīn and its rulers in the Latin East against the Muslims until 1187/ 583. Tibnīn played a key role in overcoming the Muslims in Tyre and controlled it in 1124. It also played a vital role in the conflict between Damascus and the Kingdom of Jerusalem. Tibnīn participated in defending Antioch, Banyas, Hebron and Transjordan several times. Furthermore, its soldiers and Knights joined the army of the Kingdom of Jerusalem to capture Ascalon in 1153, and joined the campaigns of Amaury I, King of Jerusalem, against Egypt from 1164 to1169. The military situation of Tibnīn under the rule of the royal house until its fall to the Muslims in 1187/ 583 will be studied as well.
Die Verwendung von photolabilen Schutzgruppen zur nicht-invasiven Kontrolle von Systemen birgt ein großes Potential für verschiedenste Anwendungsgebiete, die von der Erforschung und Regulation biologischer Prozesse, über den Einsatz in medizinischer Therapie bis hin zur Verwendung als molekulare Datenspeicher reichen. Für diese Umsetzung benötigt es allerdings eine breite Auswahl an entsprechenden PPGs und das Wissen über ihre Reaktionsmechanismen. Im Allgemeinen lässt sich die Konzeptionierung von PPGs in drei Prozesse einteilen, beginnend bei dem Design und der Synthese einer neuen PPG. Bei diesem Schritt liegt der Fokus auf ein oder zwei besonderen Eigenschaften, wie beispielsweise einer Absorptionswellenlänge in einem bestimmten Spektralbereich oder einer hohen Uncaging-Quantenausbeute. Im zweiten Schritt folgt die Untersuchung der PPG bezüglich spektroskopischer und mechanistischer Eigenschaften und ggf. anschließender Optimierung auf synthetischer Ebene. Die so gewonnenen Informationen sind dann hilfreich bei dem letzten Schritt, bei dem es um den Einsatz der PPG in einem entsprechenden System geht. Hierbei müssen die verwendeten PPGs genau auf das Zielsystem abgestimmt sein, dazu zählen verschiedenste Parameter wie Anregungswellenlänge, Extinktionskoeffizient, Art und Struktur der Photoprodukte sowie Uncaging-Effizienz und Geschwindigkeit.
In der vorliegenden Arbeit wurde über die drei vorgestellten Projekte mittels spektroskopischer Methoden zu allen drei genannten Stadien zur Konzeptionierung von PPGs ein Beitrag geleistet. Dazu zählt die Entwicklung der CBT-basierten PPGs, die Untersuchung der Struktur-Wirkungsbeziehung von (DMA)(2)F-PPGs und die Etablierung einer wellenlängenselektiven An-/Aus-Funktionalität eines Antibiotikums. In enger interdisziplinärer Zusammenarbeit zwischen theoretischen, synthetischen und biologischen Teilgebieten konnte jedes Projekt innerhalb der jeweiligen Entwicklungsstufe erfolgreich abgeschlossen werden.
Mithilfe des relativ neuen Ansatzes, bei dem durch quantenmechanische Berechnungen der vertikalen Anregungsenergie von der kationischen Spezies einer PPG-Grundstruktur eine Aussage über ihre Qualität postuliert werden kann, konnte ausgehend von der Fluoren-Grundstruktur eine neue Klasse von PPGs gefunden werden. Dabei erwies sich die CBT-Struktur mit den Schwefelatomen an der para-Position als besonders geeignet. Insbesondere konnte die Grundstruktur durch die (OMePh)2-Substitution, welche in einer signifikanten bathochromen Verschiebung des Absorptionsmaximums resultierte, optimiert werden. Die Untersuchung der Ultrakurzzeit-Dynamik beider p-CBT Strukturen gab Aufschluss über die unterschiedlichen photochemischen Eigenschaften als PPG.
Für die Stoffklasse der Dimethylamino-Fluorene wurde ein wichtiger Unterschied zwischen den einfach- und zweifach-substituierten Derivaten aufgedeckt, der entscheidend für einen signifikanten Uncaging-Effizienzunterschied ist. Dabei stellt sich die Stabilität des symmetrisch-substituierten Fluorenyl-Kations als der wichtigste Faktor bezüglich der Uncaging-Quantenausbeuten heraus. Beide Schutzgruppen sind in der Lage photoinduziert eine AG freizusetzen, wobei der Reaktionsmechanismus über die kationische Spezies (DMA)(2)F + abläuft. Der Unterschied hierbei liegt in der Lebensdauer der beiden Kationen, die im Falle der symmetrischen PPG stark lösungsmittelabhängig ist und bis zu mehreren Stunden betragen kann, was bis dato das langlebigste Kation dieser Molekülklasse darstellt. Für die zukünftige Optimierung dieser PPG-Klasse ist die Erkenntnis über die Gründe für die Stabilität des Kations von großem Vorteil. Der stabilisierende Faktor ist zum einen die zweite Dimethylamino-Gruppe der symmetrischen Verbindung, welche durch die Erweiterung der Mesomerie zur besseren Verteilung der positiven Ladung im Molekül führt. Zum anderen spielt das Lösungsmittel eine entscheidende Rolle. Dabei bieten protische, polare Medien eine zusätzliche Stabilisierung, die notwendig für die Langlebigkeit des Kations ist. Die Lebensdauer des Kations war zudem durch eine zweite Bestrahlungswellenlänge kontrollierbar. Ausgehend vom Kation konnte eine reversible Nebenreaktion in protischen Lösungsmitteln identifiziert werden, die einen Austausch der AG durch das Lösungsmittel darstellt.
Zusätzlich konnte die kleine Stoffklasse der bisher bekannten Photobasen durch die Verbindung (DMA)2F-OH erweitert werden. Genauer betrachtet handelt es sich dabei um eine photoinduzierte Hydroxidfreisetzung, wodurch je nach eingesetzter Konzentration ein pH-Sprung von bis zu drei Einheiten erreicht werden konnte. Dabei stellt sich die Lebensdauer des pH-Sprungs als ein entscheidender Parameter für Photobasen dar, welcher sich für die hier untersuchte Verbindung aufgrund der besonderen Stabilität des entsprechenden Kations, im Vergleich zu einigen der bereits bekannten Verbindungen, als besonders langlebig herausgestellt hat. Ein weiterer Vorteil des Einsatzes von (DMA)2F-OH als Photobase ist die Möglichkeit den pH-Sprung durch zwei verschiedene Wellenlängen sowohl zeitlich als auch örtlich zu kontrollieren, indem die Verbindung zwischen den zwei Spezies (DMA)2F-OH und (DMA)2F + geschaltet werden kann.
Im Hinblick auf die Anwendungen von PPGs zur verbesserten zeitlichen und örtlichen Kontrolle biologischer Zielsysteme ist im Rahmen dieser Arbeit das Prinzip vom wellenlängenselektiven Uncaging zweier PPGs an einem Molekül (two-PPG-one-molecule, TPOM) etabliert worden. Das Zielmolekül war hier das Antibiotikum Puromycin, welches durch seine Fähigkeit an das Ribosom zu binden, die Proteinbiosynthese inhibieren kann. Dabei wurden zwei verschiedene PPGs gefunden, die sowohl aufeinander als auch auf das Biomolekül selbst abgestimmt sind. Im Ausgangszustand sind beide PPGs am Puromycin angebracht, wodurch es in seiner biologischen Wirkung inaktiv ist. Befindet sich das doppelt geschützte Puromycin in der ROI, so kann es durch die Bestrahlung mit einer bestimmten Wellenlänge infolge des ersten Uncaging-Schritts aktiviert werden. Da biologische Systeme nicht statisch sind, können aktivierte Moleküle stets von der gewünschten ROI nach außen gelangen, wodurch der Anspruch der räumlichen Kontrolle nicht erfüllt wird. In diesem Fall kann durch die TPOM-Umsetzung die zweite Bestrahlungswellenlänge auf den entsprechenden Bereich angewendet werden, wodurch das Uncaging der zweiten PPG initiiert und folglich das Puromycin deaktiviert wird. Des Weiteren konnte gezeigt werden, dass die Deaktivierungswellenlänge auch in der Lage ist beide PPGs zu entfernen, wodurch eine vollständige Inaktivierung des Puromycins außerhalb der ROI garantiert werden kann.
Ist die Proteinbiosynthese längerfristig blockiert, führt das schließlich zum Zelltod. Ein großes Anwendungsgebiet dieses Antibiotikums sind die Neurowissenschaften. Aufgrund der Tatsache, dass Puromycin keine Unterscheidung zwischen eukaryotischen und prokaryotischen Zellen macht, findet es keine Anwendung in der Medizin. Eine zeitliche und örtliche Kontrolle seiner Wirkung könnte den Anwendungsbereich dieses Antibiotikums evtl. ausweiten. Das wohl naheliegendste wäre der Einsatz bei Tumorzellen, deren Behandlung durch Zytostatika auf den gesamten Körper wirken und dadurch viele schwere Nebenwirkungen verursachen.
Wie bereits weiter oben beschrieben muss für jedes Biomolekül und das entsprechende Wirkzentrum die Auswahl des passenden PPG-Paares einzeln abgestimmt werden. Dennoch lässt sich anhand des hier etablierten Systems ein Konzept für die erfolgreiche Umsetzung zukünftiger TPOM-Systeme an anderen biomolekularen Wirkstoffen zusammenfassend formulieren.
* Der erste Schritt sollte die Betrachtung des Wirkzentrums des zu modifizierenden Biomoleküls sein: Welche funktionelle Gruppe bzw. Gruppen sind entscheidend für die Bindetasche oder –stelle? Dieser Bereich des Biomoleküls soll im Zuge des Uncagings entweder blockiert oder abgespalten werden. In der unmittelbaren Nähe muss die PPG1 angebracht werden.
* Bei der Wahl von PPG1 ist das wichtigste Kriterium, dass das Biomolekül mit enthaltener Schutzgruppe in seiner Wirkung unbeeinträchtigt bleibt. Dies schränkt die Auswahl beträchtlich ein. Eine mögliche Umsetzung wäre die Anbringung einer Nitro-Gruppe falls vorhanden an einen Benzolring, welcher sich im Fall eines großen Biomoleküls in der Nähe der wichtigen funktionellen Stelle befindet.
* Die zweite PPG (PPG2), deren photoinduzierte Abspaltung zur Aktivierung des Wirkstoffs führen soll, kann strukturell frei gewählt werden. Das Auswahlkriterium hierbei ist das Absorptionsspektrum. Hierbei sollte das Absorptionsmaximum rotverschoben zur PPG1 sein, um eine unerwünschte Abspaltung zu vermeiden. Außerdem darf keine signifikante Absorption von PPG2 bei der Uncaging-Wellenlänge von PPG1 vorhanden sein.
* Beide PPGs sollten eine ähnliche Uncaging-Quantenausbeute vorweisen, um im Deaktivierungsschritt der doppelt geschützten Verbindung durch das höher energetische Licht keine Bevorzugung einer einzelnen Schutzgruppe zu riskieren.
Anhand der erarbeiteten Herangehensweise können weitere Wirkstoffe oder Biomoleküle hin zu einer An- / Aus-Funktionalität modifiziert werden. Mit der Umsetzung des TPOM-Konzepts kann eine Verbesserung der örtlichen und zeitlichen Kontrolle der Aktivität eines Antibiotikums erreicht werden. Für die Anwendung in biologischer Umgebung ist diese präzische Kontrolle essentiell, um unerwünschte Nebenwirkungen angesundem Gewebe zu verhindern.
This article discusses the counterpart of interactive machine learning, i.e., human learning while being in the loop in a human-machine collaboration. For such cases we propose the use of a Contradiction Matrix to assess the overlap and the contradictions of human and machine predictions. We show in a small-scaled user study with experts in the area of pneumology (1) that machine-learning based systems can classify X-rays with respect to diseases with a meaningful accuracy, (2) humans partly use contradictions to reconsider their initial diagnosis, and (3) that this leads to a higher overlap between human and machine diagnoses at the end of the collaboration situation. We argue that disclosure of information on diagnosis uncertainty can be beneficial to make the human expert reconsider her or his initial assessment which may ultimately result in a deliberate agreement. In the light of the observations from our project, it becomes apparent that collaborative learning in such a human-in-the-loop scenario could lead to mutual benefits for both human learning and interactive machine learning. Bearing the differences in reasoning and learning processes of humans and intelligent systems in mind, we argue that interdisciplinary research teams have the best chances at tackling this undertaking and generating valuable insights.
We focus on the role of social media as a high-frequency, unfiltered mass information transmission channel and how its use for government communication affects the aggregate stock markets. To measure this effect, we concentrate on one of the most prominent Twitter users, the 45th President of the United States, Donald J. Trump. We analyze around 1,400 of his tweets related to the US economy and classify them by topic and textual sentiment using machine learning algorithms. We investigate whether the tweets contain relevant information for financial markets, i.e. whether they affect market returns, volatility, and trading volumes. Using high-frequency data, we find that Trump’s tweets are most often a reaction to pre-existing market trends and therefore do not provide material new information that would influence prices or trading. We show that past market information can help predict Trump’s decision to tweet about the economy.
Übermäßige Gewaltanwendungen durch Polizist:innen in Deutschland sind bislang nur in Ansätzen untersucht. Das Buch liefert umfassende wissenschaftliche Befunde zu einschlägigen Situationen und ihrer strafrechtlichen Aufarbeitung. Auf Basis einer Betroffenenbefragung mit über 3.300 Teilnehmenden und über 60 qualitativen Interviews stellen sich die Fälle als komplexe Interaktionsgeschehen dar, bei deren Aufarbeitung eine besondere Definitionsmacht der Polizei sichtbar wird.
Das Forschungsprojekt KviAPol (Körperverletzung im Amt durch Polizeibeamte und -beamtinnen) untersucht polizeiliche Gewaltanwendungen, die aus Sicht der Betroffenen rechtswidrig waren, mittels quantitativer Online-Befragung sowie die polizeiliche, justizielle und zivilgesellschaftliche Perspektive auf rechtswidrige Polizeigewalt und deren Aufarbeitung in Deutschland mittels qualitativer Interviews. Zum Team gehören die Wissenschaftlichen Mitarbeiterinnen Laila Abdul-Rahman, Hannah Espín Grau und Luise Klaus sowie Prof. Dr. Tobias Singelnstein, der die Projektleitung innehat.
In relativistic heavy-ion collisions, a global spin polarization, PH, of Λ and Λ¯ hyperons along the direction of the system angular momentum was discovered and measured across a broad range of collision energies and demonstrated a trend of increasing PH with decreasing sNN−−−√. A splitting between Λ and Λ¯ polarization may be possible due to their different magnetic moments in a late-stage magnetic field sustained by the quark-gluon plasma which is formed in the collision. The results presented in this study find no significant splitting at the collision energies of sNN−−−√=19.6 and 27 GeV in the RHIC Beam Energy Scan Phase II using the STAR detector, with an upper limit of PΛ¯−PΛ<0.24% and PΛ¯−PΛ<0.35%, respectively, at a 95% confidence level. We derive an upper limit on the naïve extraction of the late-stage magnetic field of B<9.4⋅1012 T and B<1.4⋅1013 T at sNN−−−√=19.6 and 27 GeV, respectively, although more thorough derivations are needed. Differential measurements of PH were performed with respect to collision centrality, transverse momentum, and rapidity. With our current acceptance of |y|<1 and uncertainties, we observe no dependence on transverse momentum and rapidity in this analysis. These results challenge multiple existing model calculations following a variety of different assumptions which have each predicted a strong dependence on rapidity in this collision-energy range.

Elliptic flow of heavy-flavor decay electrons in Au+Au collisions at √sNN = 27 and 54.4 GeV at RHIC

We report on new measurements of elliptic flow (v2) of electrons from heavy-flavor hadron decays at mid-rapidity (|y|<0.8) in Au+Au collisions at sNN−−−√ = 27 and 54.4 GeV from the STAR experiment. Heavy-flavor decay electrons (eHF) in Au+Au collisions at sNN−−−√ = 54.4 GeV exhibit a non-zero v2 in the transverse momentum (pT) region of pT< 2 GeV/c with the magnitude comparable to that at sNN−−−√=200 GeV. The measured eHF v2 at 54.4 GeV is also consistent with the expectation of their parent charm hadron v2 following number-of-constituent-quark scaling as other light and strange flavor hadrons at this energy. These suggest that charm quarks gain significant collectivity through the evolution of the QCD medium and may reach local thermal equilibrium in Au+Au collisions at sNN−−−√=54.4 GeV. The measured eHF v2 in Au+Au collisions at sNN−−−√= 27 GeV is consistent with zero within large uncertainties. The energy dependence of v2 for different flavor particles (π,ϕ,D0/eHF) shows an indication of quark mass hierarchy in reaching thermalization in high-energy nuclear collisions.
Density fluctuations near the QCD critical point can be probed via an intermittency analysis in relativistic heavy-ion collisions. We report the first measurement of intermittency in Au+Au collisions at √sNN = 7.7-200 GeV measured by the STAR experiment at the Relativistic Heavy Ion Collider (RHIC). The scaled factorial moments of identified charged hadrons are analyzed at mid-rapidity and within the transverse momentum phase space. We observe a power-law behavior of scaled factorial moments in Au+Au collisions and a decrease in the extracted scaling exponent (ν) from peripheral to central collisions. The ν is consistent with a constant for different collisions energies in the mid-central (10-40%) collisions. Moreover, the ν in the 0-5% most central Au+Au collisions exhibits a non-monotonic energy dependence that reaches a minimum around √sNN = 27 GeV. The physics implications on the QCD phase structure are discussed.
