Methylphenidat ist ein Dopaminreuptakehemmer, der in seiner chemischen Struktur dem Amphetamin ähnlich ist. Klinisch wird es in der Behandlung des juvenilen Aufmerksamkeitsdefizit-Syndroms (ADHS) eingesetzt. Aber auch eine steigende Anzahl Erwachsener, die am ADHS leiden, profitiert von dessen therapeutischen Wirkungen. Bei gleichzeitiger Einnahme von Methylphenidat und Ethanol wird aus beiden der aktive Metabolit Ethylphenidat gebildet. Zur Charakterisierung der pharmakokinetischen Eigenschaften von Methylphenidat bei gleichzeitiger Ethanolaufnahme, wurden Untersuchungen zum in-vitro Metabolismus in humanem Leberhomogenat und ein von der Ethikkommission und der Bundesbehörde genehmigter Probandenversuch nach AMG durchgeführt. Dabei wurden drei verschiedene Konditionen mit variierter Einnahmereihenfolge der Prüfsubstanzen bei 9 gesunden männlichen Probanden untersucht, die die alleinige Aufnahme von Methylphenidat (20 mg), die Aufnahme von Methylphenidat (20 mg) 30 Minuten nach Ethanolaufnahme (Wein bis zu einer BAK von ca. 0,8 ‰) und die Einnahme von Methylphenidat (20 mg) 30 Minuten vor Ethanolaufnahme (Wein bis zu einer BAK von ca. 0,8 ‰) beinhalteten. Blutproben wurden über einen Messzeitraum bis zu 7 h entnommen und durch eine neu entwickelte validierte Methode mit Hochleistungsflüssigkeitschromatographie-Flugzeitmassenspektrometrie (LC-TOF) analysiert. Die in-vitro Versuche zeigten, dass nur in Gegenwart von Leberenzymen Ethylphenidat gebildet wurde, bei alleiniger Inkubation von Methylphenidat und Ethanol in Puffer konnte die Bildung von Ethylphenidat nicht nachgewiesen werden. In-vitro zeigten die Leberenzyme außerdem für die Ethylphenidatbildung eine Sättigung durch hohe Konzentration von Methylphenidat (Sättigung ab 0,7 mg/l) und Ethanol (Sättigung ab 5,3 g/L), die durchaus in der Anflutungsphase nach Medikamentenaufnahme in der Leber vorliegen können. Der Metabolismus zu Ritalinsäure wurde durch Ethanol deutlich gehemmt. Die enzymatische Reaktion war außerdem signifikant (p < 0,01) durch Natriumfluorid hemmbar. Bei zusätzlicher Inkubation mit Kokain (äquimolar zu Methylphenidat), konnte ebenfalls eine signifikant Verringerung der Bildung (p < 0,01) von Ethylphenidat und Ritalinsäure gezeigt werden. Dies gab einen Hinweis auf das am Metabolismus beteiligte Enzym, die humane Carboxylesterase 1A, die den Metabolismus von Kokain katalysiert. Außerdem konnte eine geringfügige Bildung von Ethylphenidat bei Inkubation von Methylphenidat und Ethanol in Serum gezeigt werden, die über eine durch Fluorid hemmbare Esterase katalysiert wurde. Im Probandenversuch fand sich in Kombination mit Ethanol ebenfalls die Bildung von Ethylphenidat. Die Konzentrationen lagen im Bereich von 0,3-3,2 mikro g/l. Methylphenidat wurde im Bereich von 4,6-28,6 mikro g/l und Ritalinsäure in einem Bereich von 187-442 mikro g/l nachgewiesen, was sich mit Ergebnissen anderer Studien deckt. Aus den quantitativen Daten wurden die pharmakokinetischen Parameter nach Einkompartimentmodell ermittelt. Für Methylphenidat wurden dabei anders als bei Kokain, bei dem sich nach Ethanolaufnahme die Wirkstoffkonzentrationen signifikant erhöhen, keine signifikanten Unterschiede bei Vergleich der 3 untersuchten Konditionen festgestellt, obwohl sich tendenziell höhere Wirkstoffkonzentrationen bei Einnahme zusammen mit Ethanol zeigten. Die Ethanolgabe vor anstatt nach Methylphenidateinnahme verzögerte die Ritalinsäurebildung (tmax 2,6 vs. 1,8 h) und im Vergleich zu der Einnahme von Methylphenidat ohne Ethanol lagen die maximalen Werte (Cmax) signifikant niedriger. Bezüglich Ethylphenidat konnte eine Erhöhung (p < 0,05) der Cmax und der Area under the curve gefunden werden, wenn Alkohol vor Methylphenidat eingenommen wurde. Im Vergleich zu Kokain, das fast 1:1 zu Kokaethylen umgesetzt wird, konnte Ethylphenidat aber nur in Spuren nachgewiesen werden. Ein Proband zeigte neben hohen Methylphenidatkonzentrationen (> 25 ng/ml) niedrige Ritalinsäurekonzentrationen (< 90 ng/ml) bei normaler Ethylphenidatbildung, was auf eine Hemmung des Methylphenidatmetabolismus bezüglich der Hydrolyse zu Ritalinsäure hindeutete. Dieser Proband wurde als „poor metabolizer“ klassifiziert. Von Kokaethylen ist bekannt, dass es bei verlängerter Halbwertszeit eine ähnlich ausgeprägte Wirkung wie Kokain besitzt. Die Eliminationshalbwertszeit von Ethylphenidat war dagegen deutlich kürzer als die von Methylphenidat (1,5 vs. 2,6 h). Die Pharmakodynamik von Ethylphenidat wurde in der Studie nicht erfasst, aus den subjektiven Berichten der Probanden ergaben sich jedoch keine deutlichen Unterschiede in der Medikamentenwirkung bei gleichzeitigem Konsum von Ethanol und Methylphenidat im Vergleich zur alleinigen Methylphenidateinnahme.
Effects of a phase transition on HBT correlations in an integrated Boltzmann+hydrodynamics approach

A systematic study of HBT radii of pions, produced in heavy ion collisions in the intermediate energy regime (SPS), from an integrated (3+1)d Boltzmann+hydrodynamics approach is presented. The calculations in this hybrid approach, incorporating an hydrodynamic stage into the Ultra-relativistic Quantum Molecular Dynamics transport model, allow for a comparison of different equations of state retaining the same initial conditions and final freeze-out. The results are also compared to the pure cascade transport model calculations in the context of the available data. Furthermore, the effect of different treatments of the hydrodynamic freeze-out procedure on the HBT radii are investigated. It is found that the HBT radii are essentially insensitive to the details of the freeze-out prescription as long as the final hadronic interactions in the cascade are taken into account. The HBT radii RL and RO and the RO/RS ratio are sensitive to the EoS that is employed during the hydrodynamic evolution. We conclude that the increased lifetime in case of a phase transition to a QGP (via a Bag Model equation of state) is not supported by the available data.

Strafe muss sein! Ein drohender Satz, eine Universalie der Gerechtigkeit, ein pädagogisches Prinzip, eine Existenzgarantie für den strafenden Staat? Wenn Strafe denn wirklich "sein muss", wie kann sie als rechtliche ausgeübt und legitimiert werden? Hinter dem Alltagsspruch "Strafe muss sein" öffnet sich das weite Feld der Geschichte des Strafrechts und der Strafrechtswissenschaft. Dieses Feld ist von der traditionellen Rechtsgeschichte eher vernachlässigt worden. Die meisten Strafrechtler halten es für verzichtbar. Vorlesungen zur Strafrechtsgeschichte gehören nicht zum Ausbildungskanon. So kamen die Innovationen in den letzten Jahren eher von außen, von der Kulturanthropologie, von der neueren Kulturgeschichte und speziell von der erfreulich entfalteten Historischen Kriminologie. Die Rechtsgeschichte hat hiervon erheblich profitiert. Sie hat ihrerseits wieder auf die historische Forschung zum Mittelalter und zur Frühen Neuzeit zurückgewirkt. Die früher manchmal beobachteten Barrieren gegenseitiger Nichtwahrnehmung scheinen nahezu verschwunden. ...
El presente ensayo resume y analiza los diferentes esfuerzos organizacionales del gobierno bolivariano de Venezuela de alcanzar una democratización de la propiedad y administración de los medios de producción. Las diferentes iniciativas y medidas adoptadas desde el 2000 – primero reforzar el cooperativismos, luego la creación de Empresas de Producción Social (EPS) y finalmente las Empresas Socialistas, como tambien el conccepto del desarrollo endógeno, orientación normativa de la organización productiva y de los programas estadales de formación laboral, son presentados y analizados con visión crítica. En respecto a la organización interna de lugares de producción se hace un análisis crítico de los modelos de cogestión, autogestión y control obrero mirando los logros y las fallas recurriendo a trabajo de campo hecho en tres fábricas: La fábrica de aluminio Alcasa; la papelera Invepal y la fábrica de válvulas Inveval. In this thesis we have studied the physics of different ultracold Bose-Fermi mixtures in optical lattices, as well as spin 1=2 fermions in a harmonic trap. To study these systems we generalized dynamical mean-field theory for a mixture of fermions and bosons, as well as for an inhomogeneous environment. Generalized dynamical mean-field theory (GDMFT) is a method that describes a mixture of fermions and bosons. This method consists of Gutzwiller mean-field for the bosons, and dynamical mean-field theory for the fermions, which are coupled on-site by the Bose-Fermi density-density interaction and possibly a Feshbach term which converts a pair of up and down fermions into a molecule, i.e. a boson. We derived the self-consistency equations and showed that this method is well-controlled in the limit of high lattice coordination number z. We develop real-space dynamical mean-field theory for studying systems in an inhomogeneous environment, e.g. in a harmonic trap. The crucial difference compared to standard DMFT is that we are taking into account that different sites are not equivalent to each other and thus take into account the inhomogeneity of the system. Different sites are coupled by the real-space Dyson equation. ...
We perform a two-flavor dynamical lattice computation of the Isgur-Wise functions t1/2 and t3/2
at zero recoil in the static limit. We find t1/2(1) = 0.297(26) and t3/2(1) = 0.528(23) fulfilling
Uraltsev’s sum rule by around 80%. We also comment on a persistent conflict between theory and
experiment regarding semileptonic decays of B mesons into orbitally excited P wave D mesons,
the so-called “1/2 versus 3/2 puzzle”, and we discuss the relevance of lattice results in this
context.
Das Ziel der vorliegenden Dissertationsarbeit ist die durchflusszytometrische Bestimmung Cytomegalievirus-spezifisch aktivierbarer CD4+T-Lymphozyten bei Patienten nach Lungentransplantation. Virusinfekte, insbesondere mit dem Cytomegalievirus (CMV), sind häufige Komplikationen nach Lungentransplantation. Bei Untersuchungen zu knochenmark-, leber-und nierentransplantierten Patienten konnte gezeigt werden, dass eine enge Korrelation zwischen der virusspezifischen, zellvermittelten T-Zellreaktion und der viralen Replikation besteht [Sesters, M. et. al., 2001 Transplantation 71, 1287-1294]. Zur Klärung der Frage, inwieweit ein Zusammenhang zwischen dem Auftreten einer CMVInfektion und dem Anteil CD4+ T-Zellen mit de-novo-Synthese von Interferon-γ (INF-γ) und CD69 bei Patienten nach einer Lungentransplantation existiert, wurde bei n=22 Patienten nach Lungentransplantation während 12 Monate der Anteil [%] virusspezifisch aktivierbarer CD4+ Lymphozyten mit de-novo-Synthese von INF-γ und CD69 gemessen. Dazu wurde peripheres venöses Blut mit CMV-KBR-Antigen inkubiert und der Anteil virusspezifisch stimulierbarer CD4+ Lymphozyten durchflusszytrometrisch quantifiziert. Bei n=7 CMV-positiven Patienten ohne akuten CMV-Infekt während des Beobachtungszeitraums (kein Nachweis von CMV-DNA und CMV-pp65) reagierten 6,13 % (median; Spannweite: 0,86-45,15%) der CD4+ T-Lymphozyten auf die Simulation mit dem Virusantigen mit einer de-novo-Synthese von INF-γ und CD69. Im Vergleich dazu zeigte sich bei n=5 CMV-positiven Patienten mit akuter CMV-Infektion im Beobachtungszeitraum ein deutlich verringerter Anteil CD4+ T-Zellen mit de-novo-Synthese von INF-γ und CD69 (0,99%; 0,01-3,20%). CMV-negative Patienten (n=10) wiesen im Vergleich zu den CMV-positiven Patienten mit und ohne CMV-Infekt im Untersuchungszeitraum einen geringeren Anteil CD4+ T-Zellen mit de-novo-Synthese von INF-γ und CD 69 (0,04%; 0,02-0,41%) auf. Bei der durchflusszytometrischen Bestimmung spezifisch aktivierbarer CD4+ T-Lymphozyten im peripheren Blut lungentransplantierter Patienten konnte ein Zusammenhang zwischen dem Anteil der virusspezifisch aktivierbaren T-Zellen und der viralen Replikation gezeigt werden. Die Methode ermöglicht eine Eingrenzung von Risikopatienten im Hinblick auf eine CMVReaktivation und kann das bestehende CMV-Monitoring sinnvoll ergänzen. Die gewonnenen Ergebnisse können Ausgangspunkt für weitere Studien zu virusspezifisch aktivierbaren T-Lymphozyten bei lungentransplantierten Patienten sein.

Revendiquer des constitutions fut l’élément dominant du xixe siècle, tenir la promesse de réaliser les droits de l’homme et la démocratie celui du xxe siècle. L’arc se tend sans peine de 1789 à 1989, et même après, il n’y a pas d’issue visible, pas d’alternative en vue. Bacterial autotransporters represent a diverse family of proteins that autonomously translocate across the inner membrane of Gram-negative bacteria via the Sec complex and across the outer bacterial membrane. They often possess exceptionally long N-terminal signal sequences. We analyzed 90 long signal sequences of bacterial autotransporters and members of the two-partner secretion pathway in silico and describe common domain organization found in 79 of these sequences. The domains are in agreement with previously published experimental data. Our algorithmic approach allows for the systematic identification of functionally different domains in long signal sequences. Background: During the last two decades the German hospital sector has been engaged in a constant process of transformation. One obvious sign of this is the growing amount of hospital privatization. To date, most research studies have focused on the effects of privatization regarding financial outcomes and quality of care, leaving important organizational issues unexplored. Yet little attention has been devoted to the effects of privatization on physicians' working routines. The aim of this observational real-time study is to deliver exact data about physicians' work at hospitals of different ownership. By analysing working hours, further impacts of hospital privatization can be assessed and areas of improvement identified.
Methods: Observations were made by shadowing 100 physicians working in private, for-profit or non-profit as well as public hospital departments individually during whole weekday shifts in urban German settings. A total of 300 days of observations were conducted. All working activities were recorded, accurate to the second, by using a mobile personal computer.
Results: Results have shown significant differences in physicians' working activities, depending on hospital ownership, concerning working hours and time spent on direct and indirect patient care.
Conclusion: This is the first real-time analysis on differences in work activities depending on hospital ownership. The study provides an objective insight into physicians' daily work routines at hospitals of different ownership, with additional information on effects of hospital privatization.
Flavins are employed to transform physical input into biological output signals. In this function, flavins catalyze a variety of light-induced reactions and redox processes. However, nature also provides flavoproteins with the ability to uncouple the mediation of signals. Such proteins are the riboflavin-binding proteins (RfBPs) with their function to store riboflavin for fast delivery of FMN and FAD. Here we present in vitro and in vivo data showing that the recently discovered archaeal dodecin is an RfBP, and we reveal that riboflavin storage is not restricted to eukaryotes. However, the function of the prokaryotic RfBP dodecin seems to be adapted to the requirement of a monocellular organism. While in eukaryotes RfBPs are involved in trafficking riboflavin, and dodecin is responsible for the flavin homeostasis of the cell. Although only 68 amino acids in length, dodecin is of high functional versatility in neutralizing riboflavin to protect the cellular environment from uncontrolled flavin reactivity. Besides the predominant ultrafast quenching of excited states, dodecin prevents light-induced riboflavin reactivity by the selective degradation of riboflavin to lumichrome. Coordinated with the high affinity for lumichrome, the directed degradation reaction is neutral to the cellular environment and provides an alternative pathway for suppressing uncontrolled riboflavin reactivity. Intriguingly, the different structural and functional properties of a homologous bacterial dodecin suggest that dodecin has different roles in different kingdoms of life.
Do we need a bad bank?

The cerebral cortex presents itself as a distributed dynamical system with the characteristics of a small world network. The neuronal correlates of cognitive and executive processes often appear to consist of the coordinated activity of large assemblies of widely distributed neurons. These features require mechanisms for the selective routing of signals across densely interconnected networks, the flexible and context dependent binding of neuronal groups into functionally coherent assemblies and the task and attention dependent integration of subsystems. In order to implement these mechanisms, it is proposed that neuronal responses should convey two orthogonal messages in parallel. They should indicate (1) the presence of the feature to which they are tuned and (2) with which other neurons (specific target cells or members of a coherent assembly) they are communicating. The first message is encoded in the discharge frequency of the neurons (rate code) and it is proposed that the second message is contained in the precise timing relationships between individual spikes of distributed neurons (temporal code). It is further proposed that these precise timing relations are established either by the timing of external events (stimulus locking) or by internal timing mechanisms. The latter are assumed to consist of an oscillatory modulation of neuronal responses in different frequency bands that cover a broad frequency range from <2 Hz (delta) to >40 Hz (gamma) and ripples. These oscillations limit the communication of cells to short temporal windows whereby the duration of these windows decreases with oscillation frequency. Thus, by varying the phase relationship between oscillating groups, networks of functionally cooperating neurons can be flexibly configurated within hard wired networks. Moreover, by synchronizing the spikes emitted by neuronal populations, the saliency of their responses can be enhanced due to the coincidence sensitivity of receiving neurons in very much the same way as can be achieved by increasing the discharge rate. Experimental evidence will be reviewed in support of the coexistence of rate and temporal codes. Evidence will also be provided that disturbances of temporal coding mechanisms are likely to be one of the pathophysiological mechanisms in schizophrenia.

The membrane-bound heterotrimeric nitrate reductase A (NarGHI) catalyzes the oxidation of quinols in the cytoplasmic membrane of Escherichia coli and reduces nitrate to nitrite in the cytoplasm. The enzyme strongly stabilizes a menasemiquinone intermediate at a quinol oxidation site (Q(D)) located in the vicinity of the distal heme b(D). Here molecular details of the interaction between the semiquinone radical and the protein environment have been provided using advanced multifrequency pulsed EPR methods. (14)N and (15)N ESEEM and HYSCORE measurements carried out at X-band ( approximately 9.7 GHz) on the wild-type enzyme or the enzyme uniformly labeled with (15)N nuclei reveal an interaction between the semiquinone and a single nitrogen nucleus. The isotropic hyperfine coupling constant A(iso)((14)N) approximately 0.8 MHz shows that it occurs via an H-bond to one of the quinone carbonyl group. Using (14)N ESEEM and HYSCORE spectroscopies at a lower frequency (S-band, approximately 3.4 GHz), the (14)N nuclear quadrupolar parameters of the interacting nitrogen nucleus (kappa = 0.49, eta = 0.50) were determined and correspond to those of a histidine N(delta), assigned to the heme b(D) ligand His-66 residue. Moreover S-band (15)N ESEEM spectra enabled us to directly measure the anisotropic part of the nitrogen hyperfine interaction (T((15)N) = 0.16 MHz). A distance of approximately 2.2 Abetween the carbonyl oxygen and the nitrogen could then be calculated. Mechanistic implications of these results are discussed in the context of the peculiar properties of the menasemiquinone intermediate stabilized at the Q(D) site of NarGHI.
We previously proposed that the dimeric cytochrome bc(1) complex exhibits half-of-the-sites reactivity for ubiquinol oxidation and rapid electron transfer between bc(1) monomers (Covian, R., Kleinschroth, T., Ludwig, B., and Trumpower, B. L. (2007) J. Biol. Chem. 282, 22289-22297). Here, we demonstrate the previously proposed half-of-the-sites reactivity and intermonomeric electron transfer by characterizing the kinetics of ubiquinol oxidation in the dimeric bc(1) complex from Paracoccus denitrificans that contains an inactivating Y147S mutation in one or both cytochrome b subunits. The enzyme with a Y147S mutation in one cytochrome b subunit was catalytically fully active, whereas the activity of the enzyme with a Y147S mutation in both cytochrome b subunits was only 10-16% of that of the enzyme with fully wild-type or heterodimeric cytochrome b subunits. Enzyme with one inactive cytochrome b subunit was also indistinguishable from the dimer with two wild-type cytochrome b subunits in rate and extent of reduction of cytochromes b and c(1) by ubiquinol under pre-steady-state conditions in the presence of antimycin. However, the enzyme with only one mutated cytochrome b subunit did not show the stimulation in the steady-state rate that was observed in the wild-type dimeric enzyme at low concentrations of antimycin, confirming that the half-of-the-sites reactivity for ubiquinol oxidation can be regulated in the wild-type dimer by binding of inhibitor to one ubiquinone reduction site.
Diese Arbeit beschäftigt sich mit der konkreten Erzeugung von 2-3D Visualisierung. Im Fokus steht der notwendige Prozess zur Erzeugung von Computergrafik.
Da die Computergrafik heut zu Tage wichtiger Bestandteil vieler Aufgabengebiete ist, sollte deren Nutzung auch allen Menschen zugänglich sein. In den vergangen Jahren blieb dies meist nur Leuten aus den Fachgebieten vorbehalten, aufgrund der Komplexität und des notwendigen „Know-how“ über die Thematik. Mittlerweile gilt diese Tatsache als überholt. Viele Erneuerung im Bereich von Hardware und Software haben es ermöglicht, dass selbst ungeübte Anwender in der Lage sind, ansehnliche 3D Grafiken an ihren PCs bei der Arbeit oder zu Hause zu erzeugen. Dies soll ebenfalls das Ziel dieser Arbeit sein. Dazu wird in eine Applikation erstellt die die Visualisierung von graphischen Primitiven unter der Verwendug von Microsofts DirectX leicht und schnell ermöglichen soll. Als Basis dient ein Rendering-Framework, welches auf einheitliches Schnittstellenkonzept setzt, um die strikte Trennung zwischen Anwender- und Fachwissen zu vollziehen.
Weitere Schwerpunkte dieser Arbeit liegen im Bereich der Modellierung von graphischen Primtiven und der Nutzung von Shadern. Dazu wird in der Modellierung der Import von archivierten Modellen umgesetzt. Die Nutzung von Shadern soll soweit vereinfacht werden, dass Anwender auf Shader beleibig zugreifen können. Dies soll durch eine Verknüpfung zwischen Shadern und Modellen erfolgen, die ebenfalls im Bereich der Modellierung erfolgt.
