Purpose: The prevalence of "ocal allergic rhinitis" within individuals suffering from perennial rhinitis remains uncertain, and patients usually are diagnosed with non-allergic rhinitis. The aim of this study was to evaluate the prevalence of a potential "local allergic rhinitis" in subjects suffering from non-allergic rhinitis in a non-selected group of young students.
Methods: 131 students (age 25.0 ± 5.1 years) with a possible allergic rhinitis and 25 non-allergic controls without rhinitis symptoms (age 22.0 ± 2.0 years) were recruited by public postings. 97 of 131 students with rhinitis were tested positive (≥3 mm) to prick testing with 17 frequent allergens at visit 1. Twenty-four 24 subjects with a house dust mite allergy, 21 subjects with a non-allergic rhinitis, and 18 non-allergic controls were further investigated at visit 2. Blood samples were taken, and nasal secretion was examined. In addition, all groups performed a nasal provocation test with house dust mite (HDM).
Results: In serum and nasal secretion, total IgE and house dust mite specific IgE significantly differed between HDM positive subjects and controls. However, no differences between non-allergic subjects and control subjects were quantifiable. Neither a nasal provocation test nor a nasal IgE to HDM allergens showed a measurable positive response in any of the non-allergic rhinitis subjects as well as the healthy controls, whilst being positive in 13 subjects with HDM allergy.
Conclusions: Nasal IgE is present in subjects with HDM allergy, but not in non-allergic rhinitis. In the investigated non-selected population, exclusive local production of IgE is absent. By implication, therefore, our findings challenge the emerging concept of local allergic rhinitis.

Populists in the EU often call for restrictions on EU immigrants’ access to welfare rights. These calls are often demagogic and parochial. This paper aims to show what exactly is both distinct and problematic with these populist calls from a normative point of view while not necessarily reducible to demagogy and parochialism. The overall aim of the paper is not to argue that all populists call for such restrictions nor to claim that all calls for such restrictions are populist. The purpose of the paper is rather humble. It only aims to show that populist calls for restrictions on EU immigrants’ access to welfare rights are characterised by two normatively problematic arguments that target two different subsets of the citizenry: what I dub for the purpose of this paper the moralists and the immoralists. It is the way populists address these two subsets of the citizenry, as well as the fact that they could simultaneously appeal to the concerns of both groups, that makes populist approaches to welfare rights both conceptually distinct to other approaches as well as potentially politically appealing to a more diverse population of voters.
Acute lymphoblastic leukemia (ALL), a neoplastic disorder of blood cells of the lymphoid lineage, is the most frequent childhood cancer. In spite of increasing survival rates, the outcome for adults, infants or relapsed patients is still less favorable, highlighting the need for novel treatment options. Reactive oxygen species (ROS) are important signaling molecules that are involved in a variety of cellular pathways. As high ROS levels lead to oxidative stress and irreversible oxidation of cellular macromolecules, the production and elimination of ROS is tightly controlled. Therefore, cells express several antioxidant molecules and enzymes, including glutathione, catalase and the thioredoxin (Trx) system, to balance ROS levels. As cancer cells were found to have increased ROS levels that could contribute to tumor progression and metastasis, they rely strongly on these antioxidant systems to prevent oxidative damage, making cancer cells especially vulnerable to ROS-inducing treatments. ROS and oxidative stress have been shown to induce programmed cell death via different pathways, however the exact mechanisms that couples oxidative signaling and cell death is not completely understood.
As a disturbance of the cellular redox homeostasis was reported during leukemia development and progression, we wanted to determine the potential of Trx inhibitors for ALL therapy. Additionally, we aimed to further understand the role of ROS and subsequent protein oxidation in the induction and execution of programmed cell death.
First, we demonstrated that the Trx1 inhibitor PX-12 induced cell death in three ALL cell lines. Further analysis of the events leading to PX-12-induced cell death in FADD-deficient (FD) Jurkat cells revealed an increase in ROS levels and oxidation-mediated dimer formation of peroxiredoxin 3 (PRDX3). Interestingly cell death was inhibited by the thiol-containing antioxidant N-acetylcysteine (NAC), but not by non-thiol-containing ROS scavengers. PX-12 treatment further induced cleavage of caspase-9 and -3 and activation of the pro-apoptotic BCL-2 protein BAK, leading us to the conclusion that mitochondria-dependent apoptosis was induced. Interestingly, we could demonstrate an important role for the BH3-only protein NOXA in the mediation of PX-12-induced apoptosis as knock-down of NOXA prevented cell death induction and BAK activation. Our findings give novel insights into the mechanism of PX-12-induced cell death in ALL cell lines and underscores the potential of PX-12 for the treatment of ALL.
To further understand the processes leading to cell death upon inhibition of the Trx system, we analyzed global protein oxidation in Jurkat FD cells upon treatment with the Trx reductase inhibitor Auranofin. In line with previous results, Auranofin induced intrinsic apoptosis that was dependent on BAK and accompanied by increased ROS levels. Using a BIAM Switch Assay followed by mass spectrometry, we demonstrated that Auranofin treatment induced oxidation of over 200 proteins. We identified several proteins whose oxidation upon Auranofin treatment was expected, like Trx1, Trx2 and several peroxiredoxins. Additionally, we verified oxidation of APAF1-interacting protein (APIP) and protein arginine N-methyltransferase (PRMT1) that are both implicated in the regulation of apoptosis. With this analysis we were able to demonstrate that Auranofin treatment leads to changes in global protein oxidation. Whether oxidation of the determined proteins changes their functionality and contributes to apoptosis induction remains to be elucidated.
As we identified BAK as an important player in PX-12- and Auranofin-induced cell death in the previous parts of this study, we wanted to further understand its involvement in ROS-mediated cell death. First analyses in wild-type (WT) and BAK-/- murine embryonic fibroblasts (MEFs) revealed that BAK was essential for Auranofin-induced cell death and that this cell death was caspase-independent in MEFs. Interestingly, BAK oxidation was induced upon treatment with Auranofin, but not upon stimulation with the apoptosis-inducing compound Etoposide. Expression of mutated BAK, with either one or both oxidation-sensitive cysteines mutated to oxidation-insensitive serines, revealed that mutating already one cysteine protected cells from Auranofin , but not Etoposide-induced cell death. Of note, mutation of the BAK BH3 domain rescued MEFs from both, Auranofin- and Etoposide-mediated cell death. The presence of cysteine residues also altered BAK interactions as observed by a mass spectrometric analysis of Auranofin-treated MEFs expressing either WT or cysteine-less BAK. We identified interactions of WT BAK with proteins involved in mitochondrial fission and vesicle transport upon Auranofin treatment. Of note, interaction with proteins involved in apoptosis, like BAX or BCL-XL, was not changed between WT and cysteine-less BAK. Our results demonstrate a critical role for BAK oxidation in Auranofin-induced cell death. Furthermore, we identified novel oxidation-dependent BAK interaction partners.
To conclude, this study highlights the potential of ROS-inducing treatments for ALL therapy and provides novel insights into the redox regulation of programmed cell death.
Ataxia-Telangiectasia (A-T), a pleiotropic chromosomal breakage syndrome, is caused by the loss of the kinase Ataxia-telangiectasia mutated (ATM). ATM is not only involved in the response to DNA damage, but also in sensing and counteracting oxidative stress. Since a disturbed redox balance has been implicated in the pathophysiology of A-T lung disease, we aimed to further explore the interplay between ATM and oxidative stress in lung cells. Using a kinetic trapping approach, we could demonstrate an interaction between the trapping mutant TRX1-CS and ATM upon oxidative stress. We could further show that combined inhibition of thioredoxin reductase (TrxR) and ATM kinase activity, using Auranofin and KU55933 respectively, induced an increase in cellular reactive oxygen species (ROS) levels and protein oxidation in lung cells. Furthermore, ATM inhibition sensitized lung cells to Auranofin-induced cell death that could be rescued by ROS scavengers. As a consequence, targeted reduction of ATM by TRX1 could serve as a regulator of oxidative ATM activation and contribute to the maintenance of the cellular redox homeostasis. These results highlight the importance of the redox-active function of ATM in preventing ROS accumulation and cell death in lung cells.
A strong decline and thinning of the Arctic sea-ice cover over the past five decades has been documented. The former multiyear sea-ice system has largely changed to an annual system and with it the dynamics of sea-ice transport across the Arctic Ocean. Less sea ice is reaching the Fram Strait and more ice and ice-transported material is released in the northern Laptev Sea and the central Arctic Ocean. This trend is expected to have a decisive impact on ice associated (“sympagic”) communities. As sympagic fauna plays an important role in transmitting carbon from the ice-water interface to the pelagic and benthic food webs, it is important to monitor its community composition under the changing environmental conditions. We investigated the taxonomic composition, abundance and distribution of sea-ice meiofauna (here heterotrophs >10 μm; eight stations) and under-ice fauna (here metazoans >300 μm; fourteen stations) in Arctic 1.5 year-old pack ice north of Svalbard. Sampling was conducted during spring 2015 by sea-ice coring and trawling with a Surface and Under-Ice Trawl. We identified 42 taxa associated with the sea ice. The total abundance of sea-ice meiofauna ranged between 580 and 17,156 ind.m–2 and was dominated by Ciliophora (46%), Copepoda nauplii (29%), and Harpacticoida (20%). In contrast to earlier studies in this region, we found no Nematoda and few flatworms in our sea-ice samples. Under-ice fauna abundance ranged between 15 and 6,785 ind.m–2 and was dominated by Appendicularia (58%), caused by exceptionally high abundance at one station. Copepoda nauplii (23%), Calanus finmarchicus (9%), and Calanus glacialis (6%) were also very abundant while sympagic Amphipoda were comparatively rare (0.35%). Both sympagic communities showed regional differences in community composition and abundance between shelf and offshore stations, but only for the under-ice fauna those differences were statistically significant. Selected environmental variables moderately explained variations in abundances of both faunas. The results of this study are consistent with predictions of diversity shifts in the new Arctic.
In der vorliegenden Untersuchung wurde das bovine, Hydroxylapatit-basierte, Knochenersatzmaterial Hypro-Oss® zunächst ex vivo überprüft, anschließend subkutan in den interskapulären Bereich von 12 weiblichen Wistar-Ratten (Testgruppe) eingebracht; bei 12 weiteren Tieren erfolgte eine Sham-Operation ohne Einbringung von Biomaterial (Kontrollgruppe). Anschließend wurde die Gewebereaktion über 30 Tage beobachtet und die Explantate jeweils nach Tag 3, 15 und 30 histologisch und histomorphometrisch untersucht.
Die histologische Analyse zeigte innerhalb des Beobachtungszeitraums von 30 Tagen eine störungsfreie Eingliederung der Hypro-Oss®-Granula in das umliegende Gewebe. Bereits 3 Tage nach Einbringung des Biomaterials waren mononukleäre Zellen erkennbar, die bis Tag 30 weiter zunahmen. Ab diesem Zeitpunkt zeigten sich auch TRAP-positive, CD-68-negative Multinukleäre Zellen, die das Ergebnis einer Fusion von Makrophagen sind und eine Fremdkörperreaktion indizierten. Nach 30 Tagen zeigten sich die Granula histologisch stabil integriert ohne Anzeichen einer immunologischen Abstoßungsreaktion.
Die CD-68-Expression der aufgefundenen Makrophagen und mehrkernigen Riesenzellen bildete ein Kriterium zur Unterscheidung der MNGCs von Osteoklasten, die ebenfalls mehrkernig sind, aber dieses Cluster of Differentiation nicht tragen. Dies charakterisiert die vorgefundenen MNGCs als Fremdkörper-Riesenzellen, da sie ebenso wie die pathologischen Riesenzellen vom Typ Langerhans CD-68 exprimieren.
Dieses Bild bestätigte sich für die Hypro-Oss®-Gruppe in der histomorphometrischen Betrachtung über eine kontinuierliche Zunahme von überwiegend CD-68-positiven Makrophagen bis zum Tag 30, während sie für die Kontrollgruppe über die gesamte Zeit rückläufig waren. Die Multinukleären Zellen erreichten dagegen bereits an Tag 15 ihren Höhepunkt, während in der Kontrollgruppe über den gesamten Beobachtungszeitraum erwartungsgemäß keine MNGCs gefunden wurden.
Der hoch signifikante Anstieg der MNGC-Zahl der Testgruppe bis Tag 15 korreliert positiv mit den Vaskularisationsdaten, was darauf hindeutet, dass die Multinukleären Zellen durch die Einbringung des Biomaterials induziert wurden und über die Sekretion des Signalmoleküls VEGF einen wesentlichen Faktor für die Blutgefäßbildung bilden.
Eine Auffälligkeit hat sich jedoch in Bezug auf das Alleinstellungsmerkmal von Hypro-Oss® gezeigt, welches bei der Aufreinigung nicht erhitzt wird. Diverse Studien haben einen Zusammenhang der Höhe der Sintertemperatur mit der Bildung von MNGCs nachgewiesen, wonach für Hypro-Oss® eine geringe Induzierung von MNGCs zu erwarten gewesen wäre als für vergleichbare, höher erhitzte bovine Knochenersatzmaterialien. Dagegen zeigten die Vaskularisationsdaten unserer Untersuchung für Hypro-Oss® im Vergleich zu 2 anderen bovinen Knochenersatzmaterialien (Bio-Oss® und BEGO OSS®) jedoch signifikant höhere Werte für die Blutgefäßbildung als dies aus der Korrelation von Sintertemperatur mit der Anzahl Multinukleärer Riesenzellen zu erwarten gewesen wäre.
Aufgrund der relativ kurzen Dauer der Beobachtung lassen sich keine belastbaren Ergebnisse in Bezug auf den zu erwartenden Materialabbau und die ossäre Integration von Hypro-Oss® feststellen, welche einer längerfristigen Analyse bedürften als es in dieser Untersuchung möglich war. Es gibt aber klinische Erfahrungsberichte23 hinsichtlich Handling, Heilungsverlauf und Materialintegration von Hypro-Oss® bei Sinusbodenelevation und Guided Bone Regeneration, die auch in der Langfristbetrachtung positive Ergebnisse zeigten. Offen bleibt, ob nicht eine physiologische Wundheilung nur mittels Makrophagen einer pathologischen Wundheilung unter Mitwirkung Multinukleärer Riesenzellen überlegen ist: zumindest robustere Knochenersatzmaterialien wie z.B. das hier untersuchte Hypro-Oss® scheinen dabei weniger sensibel auf Multinukleäre Riesenzellen zu reagieren.
Sarcomas are rare cancers with high heterogeneity in terms of type, location, and treatment. The health-related quality of life (HRQoL) of sarcoma patients has rarely been investigated and is the subject of this analysis. Adult sarcoma patients and survivors were assessed between September 2017 and February 2019 in 39 study centers in Germany using standardized, validated questionnaires (European Organization for Research and Treatment of Cancer Quality of Life Questionnaire (EORTC QLQ-C30)). Associated factors were analyzed exploratively using multivariable linear regressions. Among 1113 patients, clinically important limitations and symptoms were most pronounced in emotional (63%, 95% CI 60–66%), physical (60%, 95% CI 57–62%), role functioning (51%, 95% CI 48–54%), and pain (56%, 95% CI 53–59%) and fatigue (51%, 95% CI 48–54%). HRQoL differed between tumor locations with lower extremities performing the worst and sarcoma types with bone sarcoma types being most affected. Additionally, female gender, higher age, lower socioeconomic status, recurrent disease, not being in retirement, comorbidities, and being in treatment were associated with lower HRQoL. Sarcoma patients are severely restricted in their HRQoL, especially in functioning scales. The heterogeneity of sarcomas with regard to type and location is reflected in HRQoL outcomes. During treatment and follow-up, close attention has to be paid to the reintegration of the patients into daily life as well as to their physical abilities and emotional distress.
In this explorative study, we investigate how sequences of behaviour are related to success or failure in complex problem‐solving (CPS). To this end, we analysed log data from two different tasks of the problem‐solving assessment of the Programme for International Student Assessment 2012 study (n = 30,098 students). We first coded every interaction of students as (initial or repeated) exploration, (initial or repeated) goal‐directed behaviour, or resetting the task. We then split the data according to task successes and failures. We used full‐path sequence analysis to identify groups of students with similar behavioural patterns in the respective tasks. Double‐checking and minimalistic behaviour was associated with success in CPS, while guessing and exploring task‐irrelevant content was associated with failure. Our findings held for both tasks investigated, from two different CPS measurement frameworks. We thus gained detailed insight into the behavioural processes that are related to success and failure in CPS.
Aim: Assessment of the effect of nonsurgical periodontal therapy on haematological parameters in patients with grades B (BP) and C periodontitis (CP).
Methods: Eight BP and 46 CP patients received full-mouth periodontal debridement within 48 h, if positive for Aggregatibacter actinomycetemcomitans with adjunctive systemic antibiotics (4 BP, 17 CP). Clinical data were collected prior and 12 weeks after periodontal therapy. Blood was sampled prior to and 1 day as well as 6 and 12 weeks after the first SD visit. Erythrocyte count, haemoglobin value, haematocrit (HCT), mean erythrocyte volume (MCV), mean corpuscular haemoglobin (MCH), MCH concentration (MCHC), platelets (PLT) and heat shock protein 27 (Hsp27) were assessed.
Results: Both groups showed significant clinical improvement (p < 0.05). Using univariate analysis, MCV was noticeably lower in CP than BP at all examinations, HCT only at baseline. For CP, MCHC was noticeably higher 12 weeks after SD than at baseline and 1 day (p ≤ 0.005) and Hsp27 increased noticeably at 1 day (p < 0.05). Repeated measures analysis of variance revealed African origin to be associated with lower MCV and female sex with lower MCHC.
Conclusion: Based on multivariate analysis, periodontal diagnosis (BP/CP) was not associated with haematological parameters measured in this study or serum Hsp27. In CP, nonsurgical periodontal therapy improved MCHC 12 weeks after SD. Also in CP Hsp27 was increased 1 day after SD.
Zur Evolution der Hirnmorphologie und Anpassungen an Extremhabitate im Taxon Poecilia (Teleostei)

Diese Dissertation befasst sich mit den Auswirkungen kontrastierender Umweltbedingungen auf die Gehirnmorphologie von neotropischen Fischen der Gattung Poecilia, welche unterschiedlichen abiotischen sowie biotischen Stressoren ausgesetzt sind. Da das Gehirn der Teleostei ein energetisch kostspieliges Organ und viel plastischer ist als z. B. bei Säugetieren, stellt sich die Frage, wie die Gehirnanatomie durch divergierende ökologische Faktoren in verschiedenen Umgebungen geformt wird, die ´extreme´, ´ressourcenbeschränkte und günstige´ Umgebungen repräsentieren. Zur Beantwortung dieser Frage wurden intraspezifische Studien an freilebenden und Laborindividuen von Poecilia-Arten durchgeführt, um die evolutionäre und ökologische Formgebung des Gehirns besser verstehen zu lernen. Im ersten Teil der Arbeit wurden Gehirnvolumina verglichen zwischen reproduktiv isolierten Populationen des neotropischen Fisches Poecilia mexicana (Ntotal = 95), die in Dunkelheit leben (Cueva Luna Azufre), in einem nahegelegenen Oberflächenhabitat (El Azufre), welcher giftigen Schwefelwasserstoff enthält und einer Kombination aus beiden Stressoren Dunkelheit und H2S (Cueva del Azufre). In einer zweiten Studie wurde auf anatomische („konvergente“) Veränderungen im Teleost-Gehirn entlang eines natürlichen Gradienten von Sulfidkonzentrationen getestet. Hierfür wurden Gehirne (Ntotal = 100) von P. mexicana verglichen, die in drei Flusssystemen im Süden Mexikos unabhängig voneinander eine erhöhte Toleranz gegenüber Schwefelwasserstoff (H2S) entwickelt haben. Dazu gehörten eine phylogenetisch alte H2S-adaptierte Form (P. sulphuraria) und zwei P. mexicana Formen, welche frühere Stufen der Anpassung an H2S darstellen. Zur Überprüfung des Einflusses anderer abiotischer und biotischer Faktoren auf die Morphologie der Gehirnregionen wurde eine weitere Studie durchgeführt. Hierbei wurden die phänotypischen Variationen der Gehirnregionen und der Körpermorphologie von Poecilia vivipara-Populationen (Ntotal = 211) aus Lagunen des Restinga de Jurubatiba Nationalpark untersucht, die sich in abiotischen Umgebungsbedingungen, insbesondere in Salzgehalt, Wassertransparenz, Phosphat und Nitrat sowie biotischen Faktoren wie Prädatorendichte unterschieden. Die erste Studie zeigte lebensraumabhängige Unterschiede bei freilebenden Fischen. Bei Fischen, die in Dunkelheit ohne H2S (LA) oder in Oberflächenhabitaten mit H2S lebten, wurden vergrößerte telenzephale Lappen, kleinere Augen und optische Tekta gefunden. Fische aus der sulfidischen Höhle (CA) zeigten zusätzlich vergrößerte Corpus cerebelli. Der Vergleich mit den Gehirnen von Labor aufgezogenen weiblichen Fischen (Ntotal = 25) zeigt eine allgemeine Verringerung der Gehirngröße sowie eine geringe Abweichung der Gehirngröße zwischen Labor aufgezogenen und freilebenden Fischen. Auch in der zweiten Studie zeigten alle in H2S-haltigen Lebensräumen lebenden Fische kleinere Augen, ein kleineres optisches Tektum und ein kleineres Gehirnvolumen, jedoch größere Corpus cerebelli und Hypothalamusvolumen als Fische aus nicht-sulfidischen Lebensräumen. Flusssystem-spezifische Effekte wurden für die telenzephalen Lappen, das gesamte Gehirn und die Augengröße festgestellt, da die Geschlechter je nach Quelle des Flusssystems unterschiedlich auf das Vorhandensein von H2S reagierten. Die dritte Studie zeigt auch, dass andere Umwelteinflüsse bemerkenswerte Verschiebungen im Gehirn und in den Gehirnregionen verursachen können. Fische, die im Süßwasser leben, zeigten eine verringerte Gesamthirngröße, telenzephale Lappen, Corpus cerebelli und Hypothalamusvolumen. Darüber hinaus zeigten Fische aus Salzwasserlagunen (hypersalin), ein verringertes Volumen des optischen Tektum, während telenzephale Lappen, Corpus cerebelli und Hypothalamusvolumen im Vergleich zu Süßwasserfischen vergrößert waren. Im Brackwasser lebende Fische wiesen im Vergleich zu Süß- und Salzwasserfischen die größten Gehirnregion-Volumen auf. Darüber hinaus zeigten die Ergebnisse über die Lagunen hinweg auch Unterschiede in der Morphologie der Kopf- und Augendurchmesser. Bei Augengröße, Kopfgröße, optischem Tektum Volumen, Hypothalamusvolumen und dem Gesamthirnvolumen wurde ein sexueller Dimorphismus beobachtet. Die dargestellten Ergebnisse verdeutlichen, dass die gefundenen Muster nahezu mit denen von H2S-Fischen identisch sind. Die ausgeprägten Unterschiede in den Hirnregionen zwischen freilebenden Fischen können als Teil der Mosaikentwicklung interpretiert werden. Die Ergebnisse der Laborpopulation zeigen jedoch eine hohe phänotypische Plastizität. Diese Studie unterstreicht damit die Bedeutung der Kombination der Untersuchung von freilebenden mit im Labor lebenden Individuen zur Beantwortung von Fragen der Gehirnentwicklung. Inappropriate land management leads to soil loss with destruction of the land’s resource and sediment input into the receiving river. Part of the sediment budget of a catchment is the estimation of soil loss. In the Ruzizi catchment in the Eastern Democratic Republic of the Congo (DRC), only limited research has been conducted on soil loss mainly dealing with local observations on geomorphological forms or river load measurements; a regional quantification of soil loss is missing so far. Such quantifications can be calculated using the Universal Soil Loss Equation (USLE). It is composed of four factors: precipitation (R), soil (K), topography (LS), and vegetation cover (C). The factors can be calculated in different ways according to the characteristics of the study area. In this paper, different approaches for calculating the single factors are reviewed and validated with field work in two sub-catchments of Ruzizi River supplying the water for the reservoirs of Ruzizi I and II hydroelectric dams. It became obvious that the (R)USLE model provides the best results with revised R and LS factors. C factor calculations required to conduct a supervised classification using the Maximum Likelihood Procedure. Different C factor values were assigned to the land cover classes. The calculations resulted in a soil loss rate for the predominantly occurring Ferralsols and Leptosols of around 576 kt/yr in both catchments, when 2016 landcover and precipitation are used. This represents an area-normalized value of 40.4 t/ha/yr for Ruzizi I and 50.5 t/ha/yr for Ruzizi II due to different landcover in the two sub-catchments. The mean value for the whole study area is 47.8 t/ha/yr or even 27.1 t/ha/yr when considering land management techniques like terracing on the slopes (P factor). This work has shown that the (R)USLE model can serve as an easy to handle tool for soil loss quantification when comprehensive field work results are sparse. The model can be implemented in Geographic Information Systems (GIS) with free data; hence, a validation is crucial. It becomes apparent that the use of high resolution Sentinel 2a MSI data as the basis for C factor calculations is an appropriate method for considering heterogeneous Land Use Land Cover (LULC) patterns. In Deutschland erkranken pro Jahr bis zu 12.000 Menschen neu an Leukämie. Leukämie ist eine schwere onkologische Erkrankung, bei der reifes Knochenmarkgewebe in Folge von Mutationen unreifer und defekter Vorläuferzellen (leukämischen Blasten) verdrängt wird. Dies führt zu einer zunehmend eingeschränkten Blutbildung. Akute Leukämieformen können unbehandelt innerhalb von wenigen Wochen zum Tode führen und erfordern deshalb eine umgehende Diagnostik sowie einen raschen Therapiebeginn. Heilungschancen bestehen dann, wenn durch die Transplantation von gesunden hämatopoetischen Stammzellen (HSZT) das erkrankte Knochenmark ausreichend ersetzt wird. Leider sind Abstoßungsreaktionen des Spendermaterials (engl.: Graft-versus-Host-Disease, GvHD) keine Seltenheit.
Natürliche Killerzellen (NK-Zellen) stellen die kleinste Lymphozytenpopulation im menschlichen Blut dar und werden dem angeborenen Immunsystem zugerechnet. Sie wurden erstmals 1975 durch die Forscher Kiessling, Klein et al. entdeckt.17 Aufgrund ihrer Fähigkeit bestimmte Tumorzellen in vitro zu töten, wächst das Interesse an der Erforschung ihrer aktivierenden und inhibierenden Oberflächenrezeptoren. Die Killer-Zell-Immunoglobulin-ähnlichen Rezeptoren (KIRs) bilden dabei eine besonders diverse NKZell-Rezeptorfamilie. Lokalisiert auf Chromosom 19 liegen bis zu 17 hochpolymorphe KIRGene. Die genetische Ausstattung und Oberflächenexpression variiert von Individuum zu Individuum und bildet die Voraussetzung für die vorhandene Diversität KIR-exprimierender NK-Zellen. NK-Zellen besitzen die Fähigkeit, Gewebezellen in „körpereigen“ oder „fremd“ zu kategorisieren. Inhibitorische Killer-Immunoglobulin-ähnliche Rezeptoren (iKIR) nutzen dazu HLA-Klasse-I-Proteine (MHC-I) auf der Oberfläche gesunder Zellen. Diese schützen sie vor einem zytotoxischen NK-Zell-Angriff. NK-Zellen durchlaufen im Vorfeld einen komplexen Ausbildungssprozess48, an dessen Ende lizensierte Effektorzellen stehen. Diese können mittels gezielter Zytolyse krebstransformierte, zellulär-gestresste, sowie viralinfizierte Zellen im intakten Organismus erkennen und abtöten.
Die Spenderauswahl ist ein wichtiger Faktor für den Erfolg einer Stammzelltransplantation. Infundierte Spender NK-Zellen schützen das Transplantat, indem sie als wirksame Effektorzellen verbleibende Leukämiezellen aktiv eliminieren. Diese wünschenswerte Nebenwirkung wird als Graft-versus-Leukämie (GvL)-Effekt bezeichnet. Ruggeri et al. konnte zeigen, dass insbesondere Transplantationsstrategien, die auf KIR-Ligand-Fehlpaarungen (engl.: KIR-HLA-mismatch) basieren, zu weniger Rückfällen, weniger GvHD und einem besseren Gesamtüberleben bei Patienten mit akuter myeloischer Leukämie (AML) nach HSZT führt. Der KIR-HLA-mismatch wird mittlerweile aufgrund ausreichender Datenlage bei der Auswahl passender NK-Zell-Spender berücksichtigt und die Untersuchung auf die An- bzw. Abwesenheit bestimmter KIR-Gene (Haplotypisierung)
mittlerweile neben der HLA-Typisierung standardisiert in vielen Instituten durchgeführt. Daneben finden sich immer mehr Hinweise dafür, dass bereits einzelne allelische Polymorphismen innerhalb der KIR-Gene einzelner Spender großen Einfluss auf die Funktionalität ihrer NK-Zellen nehmen. Die allelische Subtypisierung von KIRs stellt aufgrund stetig steigender Zahlen neu entdeckter Allele eine Herausforderung dar. Im Januar 2019 sind für KIR2DL1 bereits 66 Allele beschrieben und für KIR3DL1 sogar 150 Allele in der Immuno Polymorphism Database (IPD) hinterlegt.
Die vorliegende Arbeit präsentiert ein praktikables Subtypisierungsverfahren, um allelische Unterschiede innerhalb der Genloci der NK-Zell-Rezeptoren KIR2DL1 und KIR3DL1 zu untersuchen. Für die Experimente wurden NK-Zellen von 20 gesunden Spendern funktionell untersucht und KIR-genetisch analysiert. Ziel war es innerhalb dieser Individuen besonders potente NK-Zellspender zu identifizieren und diese anhand bestimmter Polymorphismen und/ oder der Expression von KIR-Rezeptoren zu charakterisieren. Bei der Subtypisierung der KIR2DL1-Gene konnten 12 verschiedene, bereits bekannte KIR2DL1-Allele bestimmt werden. Die häufigsten Allele waren dabei 2DL1*001, *00201, *00302, *00401 und *00403. 5 der 20 Spender konnten der funktionell hochpotenten R245–Allelgruppe (AS Arginin an Pos. 245) zugeordnet werden. Spender 13 zeigte bei negativer KIR2DL1-SSP eine vermeintlich neue Nullallelvariante, Spender 20 eine neue heterozygote Variante, resultierend in der Kombination eines Arginin mit Alanin (R/A). Bei der allelischer Subtypisierung von KIR3DL1 wurden 25 verschiedene, bereits bekannte KIR3DL1-Allele bei den 20 Spendern bestimmt. Spender 2 zeigt zahlreiche, vorwiegend homozygote Abweichungen von der Referenzfrequenz, insbesondere im Exon 5, und wurde als neue Allelvariante gewertet. Die häufigsten KIR3DL1-Allele waren 3DL1*00101, *002 und *087. Mittels durchflusszytometrischer Messung konnte gezeigt werden, dass das bekannte Nullallel 3DL1*0040101 bei Spender 14 zu keiner Oberflächenexpression des Rezeptors führt215, während Spendern 11 und 16 als Träger des 3DL1*00402 Allels eine Oberflächenexpression von rund 10% präsentierten. Um die Spender NK-Zellen der gebildeten Gruppen funktionell zu testen, wurden die NK-Zellen experimentell mit vier unterschiedlichen transgenen L721.221-Zelllinien stimuliert. Die funktionelle Potenz der
gespendeten NK-Zellen wurde mittels eines CD107-Degranulationsassays gemessen. Nach aktuellem Stand sind nur für 53 der 150 KIR3DL1-Allele die allelischen Expressionsmuster untersucht worden. Dies bedeutet im Umkehrschluss, dass für rund 65% der bekannten KIR3DL1-Allele Daten zur Funktionalität fehlen, und damit der größte Anteil der ermittelten Allele von 6 Spendern der unknown-Expression (KIR3DL1u/u) Gruppe zugeordnet wurde. Die Spender 4 und 19 der high-Expressiongruppe (KIR3DL1h/h), sowie Spender 5 und 10 der KIR3DL1u/u-Gruppe mit den Allelen 3DL1*053, *087, *109 und Spender 2 als Träger zweier neuer KIR3DL1-Allele, zeigten in toto die besten funktionellen Ergebnisse in den Experimenten gegen die verwendete B-lymphoblastoide L721.221-Zellinien. Die Ergebnisse der vorliegenden Arbeit zeigen, dass bei der Spenderauswahl für NK-Zellbasierten Immuntherapie neben der Genotypisierung die allelische KIR-Subtypisierung als wertvolles Werkzeug entschiedener berücksichtigt werden sollte. Dafür ist es jedoch notwendig weiter an KIR-Subtypisierung und -Gruppierung Strategien zu arbeiten, um Natürlichen Killerzellen Wege in die klinische Standardpraxis zu bahnen.
Den photoaktivierbaren Schutzgruppen PPGs wurde als wichtige Werkzeuge, um z. B. biologische Prozesse zu untersuchen, in den letzten Jahren eine beachtliche Aufmerksamkeit zuteil. Der Einsatz von PPGs weist gegenüber chemischen Schutzgruppen wertvolle Vorteile auf, was deren Einsatz für biologische Konzepte attraktiv macht. Insbesondere, da keine weiteren Reagenzien außer Licht als Mittel für die Photolyse benötigt werden. Darüber hinaus ist es möglich, durch Einsatz moderner Lasertechnik, eine homogene Bestrahlung des Reaktionsvolumens mit einer hohen Lichtdosis auf einer, im Vergleich zu klassischen Lichtquellen, kürzeren Zeitskala zu gewährleisten.
Die Diversität der einsetzbaren photosensitiven Schutzgruppen, kommt einerseits der Vielfalt der anwachsenden biochemischen Fragestellungen insofern zugute, als dass die ausgewählten PPGs auf verschiedenste Anforderungen der zu untersuchenden Systeme zugeschnitten werden können. Anderseits kann die, durch einige Problemstellungen, erforderte chromatische Orthogonalität der eingesetzten Schutzgruppen, gewährleistet
werden, deren Umsetzung sich in den letzten Jahren in zahlreichen Studien als erfolgsversprechend erwies. Beide Aspekte sind unter anderem Gegenstand der vorliegenden Arbeit.
Zum einen sollte das Photocaged Puromycin als photolabiles Antibiotikum Derivat, mithilfe der Cumarinylmethyl-Schutzgruppe (DEACM-Puromycin) optimiert werden und mit dem vorherigen Nitrobenzyl-geschützten NVOC-Puromycin mittels spektroskopischer und biochemischer Methoden verglichen werden. Zum anderen sollte eine neue Strategie etabliert werden, mit deren Hilfe das photolytisch entschützte Puromycin erneut deaktiviert werden kann.
DEACM-Puromycin konnte mithilfe eines fünfstufigen Syntheseweges hergestellt werden. Ausgehend von 7-Amino-4-methylcumarin, dessen allylische Methylgruppe durch die Riley-Reaktion mit Selendioxid Se2O zum entsprechenden DEACM-Aldehyd oxidiert wurde und anschließende Reduzierung in Anwesenheit von NaBH4 zum Cumarinalkohol DEACM-OH. Eine nicht toxische Variante, bei welcher Selendioxid umgangen wurde, zeichnete sich ebenso als zielführend aus. DEACM-OH und Puromycin konnten im Anschluss über ein Carbonat-Intermediat miteinander als Carbamat verknüpft und mithilfe von HPLC aufgetrennt werden.
Die Vorrangigkeit des neuen photolabilen Puromycin Derivates (DEACM-Puromycin), wurde zuerst mithilfe der Laser-NMR-Spektroskopie sowie HPLC Verfahren erfasst. Spektroskopische Analysen im Rahmen einer Kollaboration mit dem AK von Professor Wachtveitl bestätigten, dass DEACM-Puromycin für biologische Anwendungen geeignetere photolytische Eigenschaften, wie z. B. einen höheren Extinktionskoeffizienten, eine bathochrome Verschiebung des Absorptionsmaximums, sowie eine höhere Quantenausbeute und Uncaging Effizienz der Photolyse, aufwies. Basierend auf einer vergleichbaren HOMO-LUMO Differenz beider Verbindungen (DEACM-OH und DEACM-Puromycin), konnte die Spaltung der Schutzgruppe mithilfe der Differenz der Fluoreszenzslebensdauern mit einer Rate von 0,71*108 s-1 charakterisiert werden. Dies war im Vergleich zum vorherigen NVOC-Puromycin um eine Größenordnung höher. Weitere durchgeführte spektroskopische Methoden wurden mittels quantenchemischer Rechnungen unterstützt, um wichtige Erkenntnisse der kinetischen und dynamischen Abläufe der Photolyse des geschützten Puromycins anzueignen, z. B.:
• Der zur Photolyse von DEACM-Puromycin konkurrierende Fluoreszenzprozess, kann durch protische Medien unterdrückt werden. Dies und die somit ermöglichten Wasserstoffbrückenbindungen, welche die entstehenden ionischen Intermediate während der Photolyse stabilisieren, könnten sich für die Erhöhung der Quantenausbeute der photolytischen Abspaltung von DEACM-Puromycin zu Nutze gemacht werden.
• Anhand von Experimenten auf der ultrakurzen Zeitskala wurde die Population eines angeregten S1-ICT-Zustandes detektiert. Bei diesem findet, in Anwesenheit von polarem Lösungsmittel, einen Ladungstransfer der Diethylaminoreste auf den Cumarinring statt.
• Polare Lösungsmittel bewirken ebenfalls den Übergang zu einem TICT Zustand, welcher als nichtstrahlende Relaxation die Fluoreszenz des DEACM-Puromycins reduziert. Die Auswahl von Substituenten sowie polaren und protischen Lösungsmitteln zur Begünstigung der ICT- sowie TICT- Zustände, könnte zukünftig zur Optimierung der Photolyse Effizienz herangezogen werden.
Die gelungene Optimierung des geschützten Puromycins als ein photosensitives Antibiotikum, durch die DEACM-Schutzgruppe, konnte mittels eines XTT-Zellviabilität-Experimentes mit sf9-Insektenzellen nachgewiesen werden. Zusätzlich konnte die lichtkontrollierte Puromycylierung zur Visualisierung neu synthetisierter Proteine als weitere Funktion des optimierten photoaktivierbaren Puromycins in Zusammenarbeit mit dem AK. Schumann (MPI für
Hirnforschung, Frankfurt am Main) nachgeprüft werden. Obwohl beide photocaged Verbindungen, DEACM- sowie das vorherige NVOC-Puromycin, eine vergleichbare Zellpermeabilität zu den präparierten Neurozellen aufwiesen, zeigte DEACM-Puromycin unter gleichen Bedingungen nach der Belichtung ein signifikant intensiveres Puromycylierungsignal als NVOC-Puromycin. Unter der Annahme, dass sich mehr NVOC- als DEACM-Puromycin in
den Zellen befand, bestätigt diese Beobachtung die Vorrangigkeit des DEACM-Derivates, aufgrund seiner bereits optimierten photochemischen Eigenschaften. Durch den Einsatz eines Zwei-Photonen-Lasers, konnte die Eignung von DEACM-Puromycin für die raumselektive Steuerung der Detektion von neu exprimierten Proteinen, mit größerer Auflösung auf subzellularem Level, nachgewiesen werden.

The respiratory chain of Escherichia coli contains two different types of terminal oxidase that are differentially regulated as a response to changing environmental conditions. These oxidoreductases catalyze the reduction of molecular oxygen to water and contribute to the proton motive force. The cytochrome bo3 oxidase (cyt bo3) acts as the primary terminal oxidase under atmospheric oxygen levels, whereas the bd‐type oxidase is most abundant under microaerobic conditions. In E. coli, both types of respiratory terminal oxidase (HCO and bd‐type) use ubiquinol‐8 as electron donor. Here, we assess the inhibitory potential of newly designed and synthesized 3‐alkylated Lawson derivatives through L‐proline‐catalyzed three‐component reductive alkylation (TCRA). The inhibitory effects of these Lawson derivatives on the terminal oxidases of E. coli (cyt bo3 and cyt bd‐I) were tested potentiometrically. Four compounds were able to reduce the oxidoreductase activity of cyt bo3 by more than 50 % without affecting the cyt bd‐I activity. Moreover, two inhibitors for both cyt bo3 and cyt bd‐I oxidase could be identified. Based on molecular‐docking simulations, we propose binding modes of the new Lawson inhibitors. The molecular fragment benzyl enhances the inhibitory potential and selectivity for cyt bo3, whereas heterocycles reduce this effect. This work extends the library of 3‐alkylated Lawson derivatives as selective inhibitors for respiratory oxidases and provides molecular probes for detailed investigations of the mechanisms of respiratory‐chain enzymes of E. coli.
A novel role for mutant mRNA degradation in triggering transcriptional adaptation to mutations

Robustness to mutations promotes organisms’ well-being and fitness. The increasing number of mutants in various model organisms, and humans, showing no obvious phenotype (Bouche and Bouchez, 2001; Chen et al., 2016b; Giaever et al., 2002; Kok et al., 2015) has renewed interest into how organisms adapt to gene loss. In the presence of deleterious mutations, genetic compensation by transcriptional upregulation of related gene(s) (also known as transcriptional adaptation) has been reported in numerous systems (El-Brolosy and Stainier, 2017; Rossi et al., 2015; Tondeleir et al., 2012); however, the molecular mechanisms underlying this response remained unclear. To investigate this phenomenon, I develop and study multiple models of transcriptional adaptation in zebrafish and mouse cell lines. I first show that transcriptional adaptation is not caused by loss of protein function, indicating that the trigger lies upstream, and find that the response involves enhanced transcription of the related gene(s). Furthermore, I observe a correlation between levels of mutant mRNA degradation and upregulation of related genes. To investigate the role of mutant mRNA degradation in triggering the response, I generate mutant alleles that do not transcribe the mutated gene and find that they fail to induce a transcriptional response and display stronger phenotypes. Transcriptome analysis of alleles displaying mutant mRNA degradation revealed upregulation of a significant proportion of genes displaying sequence similarity with the mutated gene’s mRNA, suggesting a model whereby mRNA degradation intermediates induce transcriptional adaptation via sequence similarity. Further mechanistic analyses suggested RNA-decay factors-dependent chromatin remodeling, and repression of antisense RNAs to be implicated in the response. These results identify a novel role for mutant mRNA degradation in buffering against mutations. Besides, they hold huge implications on understanding disease-causing mutations and shall help in designing mutations that lead to minimal transcriptional adaptation-induced compensation, facilitating studying gene function in model organisms.
Feasibility, design and sensitivity studies on innovative nuclear reactors that could address the issue of nuclear waste transmutation using fuels enriched in minor actinides, require high accuracy cross section data for a variety of neutron-induced reactions from thermal energies to several tens of MeV. The isotope 241Am (T1/2= 433 years) is present in high-level nuclear waste (HLW), representing about 1.8 % of the actinide mass in spent PWR UOx fuel. Its importance increases with cooling time due to additional production from the β-decay of 241Pu with a half-life of 14.3 years. The production rate of 241 Am in conventional reactors, including its further accumulation through the decay of 241Pu and its destruction through transmutation/incineration are very important parameters for the design of any recycling solution. In the present work, the 241 Am(n,f) reaction cross-section was measured using Micromegas detectors at the Experimental Area 2 of the n_TOF facility at CERN. For the measurement, the 235U(n,f) and 238U(n,f) reference reactions were used for the determination of the neutron flux. In the present work an overview of the experimental setup and the adopted data analysis techniques is given along with preliminary results.
Background: The newly introduced German pediatric screening examination at the end of the third year of life (U7a) incorporates visual function testing in particular; there is no ophthalmic screening during childhood in Germany. The purpose of this study is to investigate the relationship between participation in U7a and visual function at the preschool health examination (PHE) in the sixth year of life. Methods: This study evaluated PHE data from school enrollment years 2009/2010 to 2014/2015 of Rhineland-Palatinate, Germany. Visual acuity (VA) at PHE was assessed with Rodenstock visual acuity test device (tumbling E) wearing glasses if present. The relationship between participation in U7a and VA <0.7 at PHE was calculated for reduced monocular and binocular VA using multiple logistic regression adjusted for potential confounders. Results: Data from 189,704 children (91,041 girls) in 35 out of 36 districts were included. The first children to participate in U7a were enrolled in 2011/2012 school year. In total, 90,339 children (47.6%) had U7a before PHE, while 99,365 (52.4%) had not. VA <0.7 in at least one eye was measured at PHE in 8429 (4.4%) children, and in both eyes in 4345 (2.3%) children. Participation in U7a was not associated with VA <0.7 at PHE (odds ratio 0.99; 95% confidence interval: 0.94–1.04). Conclusions: The proportion of children with VA <0.7 at PHE was high. No beneficial effect of newly introduced German U7a pediatric screening examination was found for reduced VA at PHE.
Osteonecrosis (ON) is an acquired debilitating skeletal disorder, which is caused by a multitude of traumatic and non-traumatic etiological factors. Vascular damage, mechanical stress and increased intraosseous pressure have been discussed as contributors to ON. The optimal treatment of ON remains to be determined, since the current gold standard, core decompression, is insufficiently effective. Specific properties of mesenchymal stromal cells (MSCs) provide the rationale for their assessment in advanced stages of ON: Osteoinductive potential has been demonstrated and MSC preparations of suitable quality for use as medicinal products have been developed. Here we review the scant information on the use of allogeneic or autologous MSCs in advanced ON as well as potentially supportive data from pre-clinical studies with autologous bone marrow mononuclear cells (auto BM-MNCs), which have been studied quite extensively and the presumed therapeutic effect of which was attributed to the rare MSCs contained in these cell products. Outcomes in clinical trials with MSCs and auto-BM-MNCs remain preliminary and non-definitive, at best promising, with respect to their pharmacological effect. Clearly, though, the application of any of these cell therapies was technically feasible and safe in that it was associated with low complication rates. The heterogeneity of cell type and source, study protocols, cell manufacturing, cell properties, cell doses and surgical techniques might contribute to inconsistent results.
Der vorliegende Beitrag diskutiert, wie mithilfe von Unterrichtsvideographien die Reflexionskompetenz angehender Englischlehrkräfte bereits in der ersten Ausbildungsphase angebahnt und geschult werden kann. Er geht von der Annahme aus, dass praktizierenden Lehrkräften häufig die Gelegenheiten oder auch die Kompetenzen zur systematischen Reflexion fehlen (vgl. Kittel & Rollett, 2017) und diese bereits vorher grundgelegt werden müssen. Anhand von zwei Seminarbeispielen aus der Englischdidaktik, welche sich auf die disziplinspezifischen Heterogenitätsdimensionen Mehrsprachigkeit (vgl. u.a. Elsner & Wildemann, 2012; Niesen, 2018) und Transkulturalität (vgl. u.a. Viebrock, 2018; Kreft, 2019a, 2019b) beziehen, werden praktische Umsetzungsmöglichkeiten bzw. die wechselseitige Integration theoretischer Konzepte und unterrichtlicher Handlungen/Interaktionen illustriert. Die vorgestellten Aufgabenformate beziehen sich auf die kasuistische Fallarbeit (nach Lindow & Münch, 2014) sowie auf VierSchritt-Analysen (nach Santagata & Guarino, 2011). Als grundlegende Struktur für die Entwicklung von Reflexionskompetenz in videobasierten Lernsettings wird eine adaptierte Fassung des Modells von Aeppli und Lötscher (2016) mit den Verfahrensschritten „Erleben“, „Erkennen“, „Darstellen“, „Analysieren“ und „Alternative Szenarien entwickeln“ verwendet. Es zeigt sich, dass die gewählte Vorgehensweise Studierende in die Lage versetzt, die konzeptionelle und unterrichtspraktische Bedeutung von sprachlicher und kultureller Heterogenität im Englischunterricht zu erkennen und, in einem weiteren Schritt, Möglichkeiten zur Förderung von transkulturalitäts- bzw. mehrsprachigkeits-sensitivem Handeln zu identifizieren und somit ihre Reflexionskompetenzen zu schulen.
In der Querelle des Anciens et des Modernes ergreift Friedrich Hölderlin Partei für die Moderne. Daraus ergibt sich eine Absage an die Tragödie als höchste Kunstform und ein Bekenntnis zur Lyrik. Hölderlin gelangt jedoch zu einem ganz anderen Lyrikkonzept als etwa Hegel, der Lyrik als Artikulation von Subjektivität begreift. Für Hölderlin ist die Lyrik kein Medium subjektiver Selbstvergewisserung, sondern die vorzügliche Gattung des Erinnerns, des geduldigen Ausharrens ‘in dürftiger Zeit’ sowie des Schreibens gegen die gefährliche ‘exzentrische Begeisterung’. Vom Schreiben erhofft er sich eine Revolution der ‘Vorstellungsarten’. Hölderlin möchte der Moderne unabhängig von der ausgedienten Sprache der Tradition ein autonomes ästhetisches Profil verleihen, und so wird die Sprache der modernen Dichtung bei ihm unvermeidlich zur Sprache der obscuritas, der Dunkelheit. Hölderlins Ringen um eine genuin moderne Diktion verfremdet die überlieferte Sprache und führt zu einer Entfremdung von ihr, was wiederum Befremden auf Seiten der Rezipienten bewirkt. Hier soll zum einen der Wechsel von der Tragödie zur Lyrik als gattungspoetischem Paradigma der Moderne seit dem späten 18. Jahrhundert rekonstruiert werden. Zum anderen wird Hölderlins Verflechtung von Subjekt- und lyrischer Sprachkritik studiert, um die Spur sichtbar zu machen, die sie bis zu Adorno auslegt.

Chronische pulmonale Infektionen mit Pseudomonas aeruginosa (PA) betreffen die überwiegende Mehrheit der erwachsenen Mukoviszidose (Cystische Fibrose, CF) Patienten.
Diese Infektionen führen gesichert zu einer Abnahme der Lungenfunktion und Zunahme der Mortalität der Patienten. Atemwegsviren stehen im Verdacht pulmonale Exazerbationen bei CF-Patienten auszulösen. Unklar ist jedoch, welchen Einfluss eine chronische Infektion mit PA auf die Anfälligkeit und Reaktion des Atemwegsepithels auf virale Infektionen hat.
Das Ziel dieser Arbeit war es daher, die Interaktionen zwischen PA, humanen Rhinoviren (HRV) und primären bronchialen Epithelzellen zu untersuchen. Hierfür wurden Zellen von jeweils drei Patienten mit CF und mit Lungenemphysem aus Lungenexplantaten isoliert und in einem speziellen Air- Liquid-Interface Zellkulturmodell zu einem mukoziliär differenzierten mehrreihigen Flimmerepithel kultiviert. Chronische Infektionen wurden mit klinischen PA Isolaten für einen Gesamtzeitraum von 16 Tagen durchgeführt. Anschließend wurden die Zellen mit HRV infiziert. Schlüsselzytokine, Interferone und virale RNA wurden mittels Cytometric bead array, ELISA und qPCR bestimmt.
Rein virale Infektionen mit HRV führten zu einem Anstieg von IL-1, -6, -8, TNF- α, IP10 und IFN-b, IFN-l1 sowie ISGs und in ähnlichem Ausmaß konnte dies auch bei Coinfektionen mit einem mukoiden PA-Isolat beobachtet werden. Coinfektionen mit einem nicht-mukoiden PA-Isolat führten im Vergleich zu rein viralen Infektionen zu vermehrter Expression von IL-1β und IL-6 mRNA. Während es unter diesen Bedingungen auch auf Proteinebene zu einem Anstieg der IL-1β Konzentration kam, lag die Konzentration von freiem IL-6 Protein in nahezu allen Proben unter der Nachweisgrenze. Zellkulturmedium aus Coinfektionen mit diesem nicht-mukoiden PA-Isolat führten zudem zu einem Abbau oder einer Bindung von extern zugegebenen rekombinantem IL-6.
IL-8, IP-10, TNF-α Protein und mRNA von IFN-β, -λ1 und ISGs, sowie die Viruslast waren vergleichbar zwischen rein viralen Infektionen und bakteriell- viralen Coinfektionen. Ebenfalls keine Unterschiede wurden zwischen Zellen von Emphysem und CF-Spendern gefunden. Insgesamt zeigen diese Daten, dass eine PA-Infektion die Antwort differenzierter bronchialer Epithelzellen auf eine Virusinfektion verändern kann. Die hierdurch veränderte Immunantwort und möglicherweise eingeschränkten epithelialen Reparaturmechanismen könnten eine Ursache aggravierter viraler Infektionen in P. aeruginosa-infizierten Atemwegen darstellen.
Ein besseres Verständnis der Interaktionen zwischen chronisch-bakteriellen und viralen Atemwegsinfektionen könnte potenziell die Behandlung virus-induzierter Exazerbationen bei PA-infizierten CF-Patienten verbessern.
Professionelle Kompetenz gilt als zentrale Handlungsressource unterrichtlichen Handelns. Berufsbezogene Überzeugungen und Werte (beliefs) können dessen Qualität maßgeblich beeinflussen und sind Gegenstand zahlreicher empirischer Untersuchungen. Eine Vielzahl von Forschungsvorhaben präzisieren den gewählten Beliefbegriff hinsichtlich fachwissenschaftlicher, fachdidaktischer oder (sonder)pädagogischer Fragestellungen. In dem vorliegenden Beitrag wird zunächst unser Begriffsverständnis von professioneller Handlungskompetenz, Inklusion und Teachers’ Beliefs theoretisch begründet. Von diesen Betrachtungen ausgehend, wird das Forschungsdesign einer qualitativen Studie abgeleitet, welches die inklusionsbezogenen Überzeugungen von angehenden Lehrkräften hinsichtlich eines gemeinsamen Unterrichts beschreibt sowie Implikationen für die Hochschullehre aus Studierendenperspektive ermöglicht. Die Ergebnisse geben Aufschluss über das Heterogenitätsverständnis von Lehramtsstudierenden der Regelschule, deren vorherrschenden Rollenbildern und ideellen Überzeugungen zu inklusivem Unterricht. Sie skizzieren außerdem hochschuldidaktische Maßnahmen, welche aus Studierendenperspektive einen Beitrag für die Vorbereitung im Unterricht inklusiver Klassen leisten können. Konsequenzen, die sich aus unserer Sicht für die Ausbildung von Lehramtsstudierenden ergeben, werden mit Fokus auf die Entmystifizierung inklusionsbezogener Überzeugungen diskutiert.
Objective: To study the effect of total hip replacement (THR) on serum cartilage oligomeric matrix protein concentration (sCOMP) and its correlation with joint loading during gait in patients with unilateral hip osteoarthritis.
Design: In this prospective multimodal (clinical, biomechanical, biochemical) study blood samples from 15 patients were taken before and up to three times after THR (7 days, 3 months and 1 year), each after a resting period of at least 30 min, for analysis of sCOMP. Gait analysis was performed before and 1 year after THR to determine hip and knee joint moments.
Results: Seven days after THR, sCOMP decreased significantly compared to the preoperative measurement (p < 0.001). Three months and 1 year postoperatively, sCOMP reverted to concentrations in the range of the preoperative value. One year postoperatively, a linear correlation between sCOMP and the maximum hip flexion moment was indicated in the first half of the stance phase on the unaffected side (r = −0.736, p = 0.024). No further correlations could be determined.
Conclusions: Surprisingly, the removal of a joint affected by osteoarthritis did not have a sustained effect on sCOMP. Both before and after THR there was no scientifically substantiated correlation between sCOMP and joint moments from gait analysis. Consequently, the examination of sCOMP is not useful to detect altered joint loads that may influence degenerative changes of adjacent joints after THR.

Das Glioblastom ist der häufigste bösartige primäre Hirntumor im Erwachsenenalter. Trotz intensiver Forschungsbemühungen inklusive zahlreicher Therapiestudien liegt die mediane Gesamtüberlebenszeit selbst von Patienten in gutem Allgemeinzustand noch immer unter zwei Jahren. Deshalb ist die Erforschung möglicher Resistenzmechanismen sowie neuer therapeutischer Angriffspunkte dringend erforderlich, um effizientere Therapien zu entwickeln.
Insbesondere der Tumormetabolismus hat in den vergangenen Jahren an Bedeutung in der Krebsforschung gewonnen. Hier zeigte sich in anderen Tumorentitäten, dass der Metabolismus der Aminosäure Serin, insbesondere des Schlüsselenzyms der Serin-Biosynthese, der 3-Phosphoglyceratdehydrogenase (PHGDH), ein Ansatzpunkt für neue Therapien sein kann. Wir stellten uns die Frage, ob auch im Glioblastom der Serinstoffwechsel von Bedeutung für Wachstum und Überleben der Tumorzellen ist und ob dieser somit einen neuen therapeutischen Angriffspunkt darstellen kann.
Zur Untersuchung dieser Fragestellung wurden initial verschiedene Tumorzelllinien hinsichtlich ihrer basalen Expression von PHGDH und weiterer Schlüsselenzyme des Serinstoffwechsels auf mRNA- und Protein-Ebene analysiert. Anschließend wurde untersucht, ob sich die Expression dieser Enzyme durch Zellstress verändert. Zur weiteren Evaluierung der PHGDH als möglichem Ansatz für neue Therapien wurde deren Aktivität pharmakologisch und genetisch inhibiert. In Wachstumsanalysen wurde ferner die Abhängigkeit der Zellen von verschiedenen Serinquellen untersucht. Unter Bedingungen des Tumormikromilieus wurden Zelltod- und Redox-Stress-Parameter untersucht. Abschließend wurden die Ergebnisse durch eine PHGDH-Überexpression validiert.
Die basalen Expressionsniveaus der PHGDH unterschieden sich in den fünf analysierten Gliomzelllinien. Es zeigte sich keine Korrelation mit der Expression der
Serinhydroxymethyltransferase 1 und 2 (SHMT 1 und 2, zytoplasmatische bzw. mitochondriale Isoform), welche ebenfalls Schlüsselenzyme im Serinstoffwechsel sind. Unter Zellstress, welcher mittels Hypoxie oder Aktivierung des Nuclear factor erythroid 2-related factor 2 (Nrf2) induziert wurde, zeigte sich ein Anstieg der Expression von PHGDH und SHMT2. Im nächsten Schritt erfolgten Versuche mit CBR-5884, einem pharmakologischen Inhibitor der PHGDH. CBR-5884 reduzierte signifikant die intrazellulären Serin- und Glycin-Spiegel. Parallel zeigte sich eine dosisabhängige, signifikante Reduktion des Zellwachstums in allen getesteten Linien, ohne eine Induktion von Zelltod. Zelllinien mit niedriger PHGDH-Expression wurden zusätzlich bereits durch Entzug von Serin aus dem Kulturmedium signifikant in ihrem Wachstum gehemmt. Unter realistischen Bedingungen des Tumormikromilieus mit Sauerstoff- und Nährstoffmangel zeigte sich unter zusätzlicher PHGDH-Inhibition mittels CBR-5884 ein signifikanter Anstieg von Zelltod-Parametern und reaktiven Sauerstoffspezies (ROS) einhergehend mit einer reduzierten NADPH/NADP+-Ratio. Die Ergebnisse der pharmakologischen PHGDHInhibition konnten durch eine PHGDH-Gensuppression reproduziert werden.
Umgekehrt bestätigte eine genetische PHGDH-Überexpression den protektiven Effekt eines aktiven Serinmetabolismus für die Tumorzellen.
Zusammenfassend zeigte sich in unseren Versuchen, dass unter Bedingungen des Tumormikromilieus einige Gliomzell-Linien den Serinmetabolismus induzieren, was einen protektiven Effekt für das Überleben unter widrigen Mikromilieusbedingungen zu haben scheint. Umgekehrt reduzierte eine Inhibition der PHGDH als Schlüsselenzym der Serinsynthese unter diesen Bedingungen das Wachstum von Glioblastom-Zellen und induzierte gleichzeitig Zelltod und Redoxstress. Diese Ergebnisse rechtfertigen eine weitere präklinische Testung in in vivo Modellen. Zusammenfassend legen unsere Ergebnisse nahe, dass der Serinmetabolismus einen vielversprechenden Angriffspunkt für neue Therapiestrategien im Glioblastom darstellen könnte.
Objective: Many patients with localized prostate cancer (PCa) do not immediately undergo radical prostatectomy (RP) after biopsy confirmation. The aim of this study was to investigate the influence of “time-from-biopsy-to- prostatectomy” on adverse pathological outcomes.
Materials and Methods: Between January 2014 and December 2019, 437 patients with intermediate- and high risk PCa who underwent RP were retrospectively identified within our prospective institutional database. For the aim of our study, we focused on patients with intermediate- (n = 285) and high-risk (n = 151) PCa using D'Amico risk stratification. Endpoints were adverse pathological outcomes and proportion of nerve-sparing procedures after RP stratified by “time-from-biopsy-to-prostatectomy”: ≤3 months vs. >3 and < 6 months. Medians and interquartile ranges (IQR) were reported for continuously coded variables. The chi-square test examined the statistical significance of the differences in proportions while the Kruskal-Wallis test was used to examine differences in medians. Multivariable (ordered) logistic regressions, analyzing the impact of time between diagnosis and prostatectomy, were separately run for all relevant outcome variables (ISUP specimen, margin status, pathological stage, pathological nodal status, LVI, perineural invasion, nerve-sparing).
Results: We observed no difference between patients undergoing RP ≤3 months vs. >3 and <6 months after diagnosis for the following oncological endpoints: pT-stage, ISUP grading, probability of a positive surgical margin, probability of lymph node invasion (LNI), lymphovascular invasion (LVI), and perineural invasion (pn) in patients with intermediate- and high-risk PCa. Likewise, the rates of nerve sparing procedures were 84.3 vs. 87.4% (p = 0.778) and 61.0% vs. 78.8% (p = 0.211), for intermediate- and high-risk PCa patients undergoing surgery after ≤3 months vs. >3 and <6 months, respectively. In multivariable adjusted analyses, a time to surgery >3 months did not significantly worsen any of the outcome variables in patients with intermediate- or high-risk PCa (all p > 0.05).
Conclusion: A “time-from-biopsy-to-prostatectomy” of >3 and <6 months is neither associated with adverse pathological outcomes nor poorer chances of nerve sparing RP in intermediate- and high-risk PCa patients.
In the present paper, we tested the ability of individuals to judge correctly whether athletes are lying or telling the truth. For this purpose, we first generated 28 videos as stimulus material: in half of the videos, soccer players were telling the truth, while in the other half, the same soccer players were lying. Next, we tested the validity of these video clips by asking N = 65 individuals in a laboratory experiment (Study 1a) and N = 52 individuals in an online experiment (Study 1b) to rate the level of veracity of each video clip. Results suggest that participants can distinguish between true and false statements, but only for some clips and not for others, indicating that some players were better at deceiving than others. In Study 2, participants again had to make veracity estimations, but we manipulated the level of information given, as participants (N = 145) were randomly assigned to one of three conditions (regular video clips, mute video clips, and only the audio stream of each statement). The results revealed that participants from the mute condition were less accurate in their veracity ratings. The theoretical and practical implications of these findings are discussed.
Aims: Preventing hospitalization by detecting early evidence of heart failure (HF) decompensation in an outpatient setting can improve patient's quality of life and reduce costs of care. The purpose of this study was to assess the value of cardiac acoustic biomarkers (CABs), a combination of cardiohaemic vibrations synchronized with ECG signals, and heart rate (HR) for detecting HF decompensation during first 3 months after hospital discharge for HF.
Methods and results: Patients with an ejection fraction ≤35% (HFrEF) and hospitalized for decompensated HF were enrolled in a prospective observational study. All subjects wore a wearable cardioverter‐defibrillator (ZOLL LifeVest®, Pittsburgh, PA, USA) that is capable of recording CABs and HR. The primary endpoint of the study was the first HF event, defined as HF readmission or HF emergency room visit. From June 2017 through August 2019, 671 patients with HFrEF were enrolled. Eighty‐one patients (12.1%) had a total of 112 HF events. The algorithm detected HF events with a median of 32 days (interquartile range = 11‐45) in advance of the first HF event. The algorithm had a sensitivity of 69%, specificity of 60%, positive predictive value of 19%, and a negative predictive value of 94%. Of note, the baseline (first 7 days post‐enrolment) algorithm using CABs and HR was superior to New York Heart Association classification in detecting patients more likely to have HF decompensation (sensitivity and specificity of 61% and 68% vs. 46% and 55%, respectively).
Conclusions: This prospective international registry showed that an algorithm incorporating CABs and HR data detected HF events 30 days in advance of the event in patients with HFrEF during first 3 months after hospital discharge. Therefore, integrating CAB technology into clinical practice may prevent HF rehospitalizations.
Für Patienten mit lebensbedrohlichen Herzkrankheiten ist die Extracorporeal-Life-Support-Behandlung (ECLS) eine sinnvolle Therapiemöglichkeit. Sie bietet für Patienten im kardiogenen Schock ein Zeitfenster, um eine myokardiale Erholung zu erreichen. Hierbei kann in Abhängigkeit vom Krankheitsbild die zusätzliche Anwendung von IABP die Heilungschancen begünstigen. In der durchgeführten retrospektiven Studie wurden 118 Patienten betrachtet, die in der Klinik für Thorax-, Herz- und thorakale Gefäßchirurgie der Universität Frankfurt am Main im Zeitraum von Dezember 2001 bis Ende 2013 eine ECLS-Therapie erhalten haben. Bei 59 Patienten wurde die ECLS-Unterstützung in Kombination mit IABP durchgeführt. Die beiden Patientenkollektive - mit und
ohne IABP- sind hinsichtlich ihrer Risikofaktoren vergleichbar. Ausgehend von der Zielsetzung dieser Arbeit wurde analysiert, ob der gleichzeitige Einsatz der IABP bei ECLS – Therapie von Vor- oder sogar von Nachteil ist. Hierfür wurden für die beiden Therapiegruppen Überlebenszeitanalysen nach Kaplan-Meier durchgeführt. Der statistische Vergleich der Überlebensraten und des Weaningerfolgs erfolgte mit Hilfe des Log-Rank-Tests.
Die Auswertung der erhobenen Daten hat ergeben, dass kein signifikanter Unterschied bei der 30-Tages-Überlebensrate und dem Weaningerfolg für die beiden Patientenkollektive mit und ohne zusätzliche IABP-Anwendung vorhanden ist. Risikofaktoren wie ein hohes Lebensalter oder eine bereits vor Einlieferung stattgefundene Intubation verringern außerdem die Überlebenschancen nach ECLS - Therapie. Ein fortgeschrittenes NYHA-Stadium konnte nicht als negativer prädiktiver Faktor identifiziert werden.
In der Literatur kommt man bezüglich der Mortalität unter alleiniger ECLS-Therapie oder dem zusätzlichen Einsatz einer IABP zu unterschiedlichen Ergebnissen. Diese besagen teilweise, dass ECLS und IABP einander ergänzende Methoden sind, die sich durchaus synergistisch auf den Behandlungserfolg auswirken können und dass die Mortalität beim zusätzlichen Einsatz einer IABP signifikant niedriger ist. Die Durchführung weiterer prospektiver Studien mit vergleichbaren Patientenkollektiven zur Untersuchung des Outcomes bei den unterschiedlichen Behandlungsmethoden ist jedoch erforderlich, um ein aussagekräftiges Fazit ziehen zu können.
Die Qualifizierung von Sportlehrkräften für einen inklusiven Sportunterricht stellt eine zentrale Herausforderung für die sportdidaktische Lehre und Forschung dar. Obwohl die Bedeutung situationsspezifischer Fähigkeiten für die universitäre Sportlehrkräftebildung auch im Zusammenhang mit einem inklusiven Sportunterricht betont wird, fehlt es an Kompetenzmodellen, die die Ebene der situationsspezifischen Fähigkeiten berücksichtigen und somit eine Grundlage für Lehrkonzepte bilden können, die auf die Förderung situationsspezifischer Fähigkeiten für einen inklusiven Sportunterricht abzielen. Zudem mangelt es bestehenden Lehrkonzepten auch an Arbeits- und Organisationsformen, um gezielt situationsspezifische Fähigkeiten für einen inklusiven Sportunterricht anzusprechen. Schließlich existieren bisher keine Testverfahren, mit denen situationsspezifische Fähigkeiten für einen inklusiven Sportunterricht gemessen und darauf abzielende Lehrkonzepte evaluiert werden können. Ziel des vorliegenden Beitrags ist es demzufolge, konzeptionelle Überlegungen anzustellen, wie ein Kompetenzmodell für den inklusiven Sportunterricht generiert werden kann, welches die Ebene der situationsspezifischen Fähigkeiten berücksichtigt, wie ein Lehrformat angelegt sein müsste, um gezielt die situationsspezifischen Fähigkeiten für einen inklusiven Sportunterricht anzusprechen und wie diese situationsspezifischen Fähigkeiten gemessen werden können, um eine diesbezügliche Evaluation zu ermöglichen.
This afterword addresses the complex temporal and global dynamics of the coronavirus pandemic. After considering some of the new social rhythms that have emerged in the wake of Covid-19 around the world, it turns to the role of collective memory before, during and after corona. The aim is to provide a basic grid for how the Covid-19 pandemic could be addressed using memory studies expertise and concepts such as premediation, memorability, memory (ab)use, national memory, colonial memory, racial stereotypes, the digital archive, generational memory, or Anthropocene time.

Selective sympathetic and parasympathetic pathways that act on target organs represent the terminal actors in the neurobiology of homeostasis and often become compromised during a range of neurodegenerative and traumatic disorders. Here, we delineate several neurotransmitter and neuromodulator phenotypes found in diverse parasympathetic and sympathetic ganglia in humans and rodent species. The comparative approach reveals evolutionarily conserved and non-conserved phenotypic marker constellations. A developmental analysis examining the acquisition of selected neurotransmitter properties has provided a detailed, but still incomplete, understanding of the origins of a set of noradrenergic and cholinergic sympathetic neuron populations, found in the cervical and trunk region. A corresponding analysis examining cholinergic and nitrergic parasympathetic neurons in the head, and a range of pelvic neuron populations, with noradrenergic, cholinergic, nitrergic, and mixed transmitter phenotypes, remains open. Of particular interest are the molecular mechanisms and nuclear processes that are responsible for the correlated expression of the various genes required to achieve the noradrenergic phenotype, the segregation of cholinergic locus gene expression, and the regulation of genes that are necessary to generate a nitrergic phenotype. Unraveling the neuron population-specific expression of adhesion molecules, which are involved in axonal outgrowth, pathway selection, and synaptic organization, will advance the study of target-selective autonomic pathway generation.
Geochemical analyses result of prehistoric pottery from the site of Tol-e Kamin (Fars, Iran) by pXRF

A series of pottery samples from the Iranian site Tol-e Kamin, ranging from pre-historical period to the New Elamite, were analyzed in order to study the geochemical variability of the pottery assemblage. A total amount of 168 measurements were obtained using a portable XRF device and were statistically handled. The results could successfully distinguish the geochemical composition of potteries from the chalcolithic to the New Elamite periods in the Kur River Basin. A major shift in the use of different clay sources could be detected since the Proto Elamite period and afterward, in which the carbonated and marl content clays represented by Ca, Ba and Sr shifted to clay sources with a tendency to non-carbonate silty clay Al, Ti, and Fe from a different geological background. The results stress the importance of further provenance studies to address issues of trade and exchange possibilities in southwestern Iran.
This paper examines and rejects two normative justifications for low-fee private schools (LFPS), whose expansion throughout the Global South in recent years has been significant. The first justification – what I shall call the ideal thesis – contends that LFPS are the best mechanism to expand access to quality education, particularly at the primary level, and that the premise of their success is that they reject educational equality and state intervention in educational affairs, traditionally associated with public schools, embracing instead educational adequacy and unregulated markets for education. Against this thesis, the paper argues that an ideal educational arrangement must not do away with educational equality and some degree of state interference. The other justification for LFPS – the secondbest thesis – contends that although LFPS do not represent the ideal state of affairs, they nonetheless bring us a step closer to the ideal of universal primary education; they are a ‘realistic’ approximation to that goal. Against the second-best thesis, the paper argues that this justification commits the approximation fallacy: by deviating from the ideal educational arrangement LFPS may obstruct rather than facilitate its achievement.
Les trois auteurs et voyageurs étudiés dans cet article (Albert Londres, André Gide, Michel Leiris) ont prêté leurs voix à l’anticolonialisme. Ils ont critiqué les effets du colonialisme européen en Afrique noire de la fin des années 1920 au début des années 1930. Toutefois, pour des raisons diverses allant d’aspects institutionnels aux sensibilités personnelles, leur engagement ne paraissait pas tout à fait le bienvenu, d’où leur hésitation à placer la parole politique au premier plan et à assumer le rôle d’intellectuel engagé. Ils ont alors justifié leur démarche par l’expérience personnelle et singulière du voyage et par l’urgence du sujet de leurs récits. Les formes littéraires qui en résultent (grand reportage, journal intime de voyage, journal ethnographique) ont profondément modernisé le genre du récit de voyage dans un moment critique de son évolution.
Determination of a minimal postmortem interval via age estimation of necrophagous diptera has been restricted to the juvenile stages and the time until emergence of the adult fly, i.e. up until 2–6 weeks depending on species and temperature. Age estimation of adult flies could extend this period by adding the age of the fly to the time needed for complete development. In this context pteridines are promising metabolites, as they accumulate in the eyes of flies with increasing age. We studied adults of the blow fly Lucilia sericata at constant temperatures of 16 °C and 25 °C up to an age of 25 days and estimated their pteridine levels by fluorescence spectroscopy. Age was given in accumulated degree days (ADD) across temperatures. Additionally, a mock case was set up to test the applicability of the method. Pteridine increases logarithmically with increasing ADD, but after 70–80 ADD the increase slows down and the curve approaches a maximum. Sex had a significant impact (p < 4.09 × 10−6) on pteridine fluorescence level, while body-size and head-width did not. The mock case demonstrated that a slight overestimation of the real age (in ADD) only occurred in two out of 30 samples. Age determination of L. sericata on the basis of pteridine levels seems to be limited to an age of about 70 ADD, but depending on the ambient temperature this could cover an extra amount of time of about 5–7 days after completion of the metamorphosis.
Die Stichtagserhebung der Kriminologischen Zentralstelle (KrimZ) fragt jedes Jahr zum Stichtag am 31. März die Gegebenheiten in allen sozialtherapeutischen Einrichtungen deutschlandweit ab. Inzwischen liegen Daten aus 23 Erhebungsjahren vor und geben Aufschluss über die Entwicklungen der Versorgungslage (Anzahl der Einrichtungen bzw. Haftplätze), bezüglich der demografischen Daten der Gefangenen (Alter, Staatsbürgerschaft, Dauer der Haftstrafe, schwerste Straftat, Vorstrafen), über institutionelle Vorgänge (Aufnahmen, Abgänge und Nachbetreuung) sowie hinsichtlich von Daten zum Personal (Anzahl der Personalstellen und Frauenanteil). Die vorliegenden Auswertungen verdeutlichen die Entwicklungstrends in der Sozialtherapie zwischen 1997 und 2019 und legen nahe, dass nach einem starken Ausbau der sozialtherapeutischen Einrichtungen ab 1969 nun mit 71 Einrichtungen eine Sättigungsgrenze erreicht zu sein scheint. Die inhaftierten Personen werden zunehmend älter, sodass 2019 die über 50-Jährigen die größte Altersgruppe stellen. Schon seit 2003 liegt der Anteil derjenigen, die aufgrund eines Sexualdelikts inhaftiert sind, bei ca. 50 %, was gegenüber anderen Deliktgruppen eine deutliche Mehrheit darstellt. Ein Großteil der Gefangenen hat keine Haftlockerungen, wobei hier eine zunehmend restriktivere Praxis zu erkennen ist. Die Personalausstattung hat sich über die letzten 23 Jahre insofern verändert, als dass mehr Fachdienste und tendenziell weniger Stellen im allgemeinen Vollzugsdienst (AVD) eingerichtet wurden.
BH3 mimetics are novel anticancer therapeutics that induce apoptosis by targeting anti‐apoptotic BCL‐2 proteins. Highly specific inhibitors of the main anti-apoptotic proteins BCL-2, BCL‐XL and MCL‐1 promise new opportunities for the treatment of AML. However, it is currently unclear which of these anti-apoptotic BCL-2 proteins represents the most promising target in AML. Therefore, we investigated the effect of BH3 mimetics targeting either BCL-2 (ABT-199, S55746), BCL-XL (A-1331852) or MCL-1 (S63845) on eleven AML cell lines. Drug sensitivity screening revealed heterogeneous sensitivity towards the different BH3 mimetics, with the best responses observed upon targeting of MCL-1. Selected cell lines that displayed sensitivity towards the specific BH3 mimetics underwent intrinsic apoptosis, which was characterized by loss of mitochondrial membrane potential, exposure of phosphatidylserine and activation of caspases. Furthermore, S63845 turned out to displace BIMS and NOXA from MCL-1 to induce apoptotic cell death. Importantly, the translational relevance of this study was demonstrated by experiments in primary AML blasts, which displayed similar sensitivity towards BH3 mimetics as the cell lines did. Additionally, experiments with nonmalignant cells could confirm the clinical relevance of the MCL-1 inhibitor. There we could show, that S63845 does not cause cytotoxicity on HPCs at efficacious doses.
In conclusion, our findings reveal that the inhibition of BCL-2 proteins, especially MCL-1, by BH3 mimetics can be a promising strategy in AML treatment.
A tale of two seasons: The link between seasonal migration and climatic niches in passerine birds

The question of whether migratory birds track a specific climatic niche by seasonal movements has important implications for understanding the evolution of migration, the factors affecting species' distributions, and the responses of migrants to climate change. Despite much research, previous studies of bird migration have produced mixed results. However, whether migrants track climate is only one half of the question, the other being why residents remain in the same geographic range year-round. We provide a literature overview and test the hypothesis of seasonal niche tracking by evaluating seasonal climatic niche overlap across 437 migratory and resident species from eight clades of passerine birds. Seasonal climatic niches were based on a new global dataset of breeding and nonbreeding ranges. Overlap between climatic niches was quantified using ordination methods. We compared niche overlap of migratory species to two null expectations, (a) a scenario in which they do not migrate and (b) in comparison with the overlap experienced by closely related resident species, while controlling for breeding location and range size. Partly in accordance with the hypothesis of niche tracking, we found that the overlap of breeding versus nonbreeding climatic conditions in migratory species was greater than the overlap they would experience if they did not migrate. However, this was only true for migrants breeding outside the tropics and only relative to the overlap species would experience if they stayed in the breeding range year-round. In contrast to the hypothesis of niche tracking, migratory species experienced lower seasonal climatic niche overlap than resident species, with significant differences between tropical and nontropical species. Our study suggests that in seasonal nontropical environments migration away from the breeding range may serve to avoid seasonally harsh climate; however, different factors may drive seasonal movements in the climatically more stable tropical regions.
This article discusses the potential of a historical approach to sustainability transformations. Using environmental issues and governance structures as case studies, it first describes how historical “sustainability transformations” can be conceptualized. It then suggests that 19th-century constitutional reforms can be read as attempts at reaching fiscal sustainability, whereas some social reforms can be interpreted as attempts to render the capitalist economy sustainable. In conclusion, the article highlights that the primary value of historical approaches to sustainability transformations will not lie in models, but in encouraging more creative questions.
This paper presents causal evidence of the effects of boardroom networks on firm value. We exploit exogenous variation in network centrality arising from a ban on interlocking directorates of Italian financial and insurance companies. We leverage this shock to show that firms that become more central in the network as a result of the shock experience positive abnormal returns around the announcement date. We find that information dissemination plays a central role: results are driven by firms that have higher idiosyncratic volatility, low analyst coverage, and more uncertainty surrounding their earnings forecasts. We also find that firms benefit more from boardroom centrality when they are more central in the input-output network, as this reinforces information complementarities, or when they are less central in the cross-ownership network, as well as when they suffer from low profitability and low growth opportunities. Network centrality also results in higher compensation for board directors.
The turnover time of terrestrial ecosystem carbon is an emergent ecosystem property that quantifies the strength of land surface on the global carbon cycle–climate feedback. However, observation- and modeling-based estimates of carbon turnover and its response to climate are still characterized by large uncertainties. In this study, by assessing the apparent whole ecosystem carbon turnover times (τ) as the ratio between carbon stocks and fluxes, we provide an update of this ecosystem level diagnostic and its associated uncertainties in high spatial resolution (0.083∘) using multiple, state-of-the-art, observation-based datasets of soil organic carbon stock (Csoil), vegetation biomass (Cveg) and gross primary productivity (GPP). Using this new ensemble of data, we estimated the global median τ to be 43+7−7 yr (median+difference to percentile 75−difference to percentile 25) when the full soil is considered, in contrast to limiting it to 1 m depth. Only considering the top 1 m of soil carbon in circumpolar regions (assuming maximum active layer depth is up to 1 m) yields a global median τ of 37+3−6 yr, which is longer than the previous estimates of 23+7−4 yr (Carvalhais et al., 2014). We show that the difference is mostly attributed to changes in global Csoil estimates. Csoil accounts for approximately 84 % of the total uncertainty in global τ estimates; GPP also contributes significantly (15 %), whereas Cveg contributes only marginally (less than 1 %) to the total uncertainty. The high uncertainty in Csoil is reflected in the large range across state-of-the-art data products, in which full-depth Csoil spans between 3362 and 4792 PgC. The uncertainty is especially high in circumpolar regions with an uncertainty of 50 % and a low spatial correlation between the different datasets (0.2<r<0.5) when compared to other regions (0.6<r<0.8). These uncertainties cast a shadow on current global estimates of τ in circumpolar regions, for which further geographical representativeness and clarification on variations in Csoil with soil depth are needed. Different GPP estimates contribute significantly to the uncertainties of τ mainly in semiarid and arid regions, whereas Cveg causes the uncertainties of τ in the subtropics and tropics. In spite of the large uncertainties, our findings reveal that the latitudinal gradients of τ are consistent across different datasets and soil depths. The current results show a strong ensemble agreement on the negative correlation between τ and temperature along latitude that is stronger in temperate zones (30–60∘ N) than in the subtropical and tropical zones (30∘ S–30∘ N). Additionally, while the strength of the τ–precipitation correlation was dependent on the Csoil data source, the latitudinal gradients also agree among different ensemble members. Overall, and despite the large variation in τ, we identified robust features in the spatial patterns of τ that emerge beyond the differences stemming from the data-driven estimates of Csoil, Cveg and GPP. These robust patterns, and associated uncertainties, can be used to infer τ–climate relationships and for constraining contemporaneous behavior of Earth system models (ESMs), which could contribute to uncertainty reductions in future projections of the carbon cycle–climate feedback. The turnover time of terrestrial carbon (τ) controls the global carbon cycle – climate feedback and, yet, is poorly simulated by the current Earth System Models (ESMs). In this study, by assessing apparent carbon turnover time as the ratio between carbon stocks and fluxes, we provide a new, updated ensemble of diagnostic terrestrial carbon turnover times and associated uncertainties on a global scale using multiple, state-of-the-art, observation-based datasets of soil organic carbon stock (Csoil), vegetation biomass (Cveg) and gross primary productivity (GPP). Using this new ensemble, we estimated the global average τ to be 42$% &' years when the full soil depth is considered, longer than the previous estimates of 23$) &* years. Only considering the top 1 m (assuming maximum active layer depth is up to 1 meter) of soil carbon in circumpolar regions yields a global τ of 35$) &' years. Csoil in circumpolar regions account for two thirds of the total uncertainty in global τ estimates, whereas Csoil in non-circumpolar contributes merely 9.38%. GPP (2.25%) and Cveg (0.05%) contribute even less to the total uncertainty. Therefore, the high uncertainty in Csoil is the main factor behind the uncertainty in global τ, as reflected in the larger range of full-depth Csoil (3152-4372 PgC). The uncertainty is especially high in circumpolar regions with a behaviour of ESMs which could contribute to uncertainty reductions in future projections of the carbon cycle - climate feedback. Overall, we argue that current global datasets do not support robust estimates of τ globally, for which we need clarification on variations of Csoil with soil depth and stronger estimates of Csoil in circumpolar regions. Despite the large variation in both magnitude and spatial patterns of τ, we identified robust features in the spatial patterns of τ that emerge regardless of soil depth and differences in data sources of Csoil, Cveg and GPP. Our findings show that the latitudinal gradients of τ are consistent across different datasets and soil depth. Furthermore, there is a strong consensus on the negative correlation between τ and temperature along latitude that is stronger in temperate zones (30ºN-60ºN) than in subtropical and tropical zones (30ºS30ºN). The identified robust patterns can be used to infer the response of τ to climate and for constraining contemporaneous behaviour of ESMs which could contribute to uncertainty reductions in future projections of the carbon cycle - climate feedback. Sphingosine‐1‐phosphate lyase 1 (S1P lyase or SGPL1) is an essential sphingosine‐1‐phosphate‐degrading enzyme. Its manipulation favors onset and progression of colorectal cancer and others in vivo. Thus, SGPL1 is an important modulator of cancer initiation. However, in established cancer, the impact of retrospective SGPL1 modulation is elusive. Herein, we analyzed how SGPL1 siRNA affects malignancy of the human colorectal cancer cells DLD‐1 and found that in parallel to the reduction of SGPL1 expression levels, migration, invasion, and differentiation status changed. Diminished SGPL1 expression was accompanied with reduced cell migration and cell invasion in scratch assays and transwell assays, whereas metabolic activity and proliferation was not altered. Decreased migration was attended by increased cell–cell‐adhesion through upregulation of E‐cadherin and formation of cadherin‐actin complexes. Spreading cell islets showed lower vimentin abundance in border cells. Furthermore, SGPL1 siRNA treatment induced expression of epithelial cell differentiation markers, such as intestinal alkaline phosphatase and cytokeratin 20. Hence, interference with SGPL1 expression augmented a partial redifferentiation of colorectal cancer cells toward normal colon epithelial cells. Our investigation showed that SGPL1 siRNA influenced tumorigenic activity of established colorectal cancer cells. We therefore suggest SGPL1 as a target for lowering malignant potential of already existing cancer.
In this paper we adapt the Hamiltonian Monte Carlo (HMC) estimator to DSGE models, a method presently used in various fields due to its superior sampling and diagnostic properties. We implement it into a state-of-theart, freely available high-performance software package, STAN. We estimate a small scale textbook New-Keynesian model and the Smets-Wouters model using US data. Our results and sampling diagnostics confirm the parameter estimates available in existing literature. In addition, we find bimodality in the Smets-Wouters model even if we estimate the model using the original tight priors. Finally, we combine the HMC framework with the Sequential Monte Carlo (SMC) algorithm to create a powerful tool which permits the estimation of DSGE models with ill-behaved posterior densities.

In der Frankfurter Schirn sind die Gäste aufgefordert, kreativ zu werden: Für das Projekt “Today I feel like…” hat der Künstler Jeppe Hein die sonst weißen Mauern des Ausstellungshauses mit blauen Ovalen versehen, in denen die Besucherinnen und Besucher ihrer eignen Gefühlswelt Ausdruck verleihen können.
Chiralität ist in der belebten Natur ein omnipräsentes Phänomen und beschreibt die Symmetrieeigenschaft eines Objektes, dass dieses von seinem Spiegelbild unterscheidbar ist. Die bisherigen Untersuchungen der Wechselwirkung zwischen chiralen Molekülen und Licht fokussieren sich auf das Regime der Ein- und Multiphoton-Ionisation und wird mit dieser Arbeit um Untersuchungen im Starkfeldregime erweitert. Im Rahmen dieser Arbeit wurden Experimente an einzelnen chiralen Molekülen in starken Laserfeldern vorbereitet, durchgeführt, analysiert und alle geladenen Fragmente in Koinzidenz untersucht.
Die Präsentation der Ergebnisse orientierte sich an der Reihenfolge, in der auch die Datenauswertung von Vielteilchenaufbrüchen vonstattengeht: Zunächst wurde der Dichroismus in den Photoionen (PICD) auf chirale Signale in integraler differentieller Form untersucht, dann wurde die Asymmetrien in den Elektronenverteilungen vorgestellt und abschließend die Zusammenhänge zwischen den Ionen- und Elektronenverteilungen aufgezeigt.
Kapitel 6 untersuchte die (differentielle) Ionisations- und Fragmentationswahrscheinlichkeit von verschiedenen chiralen Molekülen. Die in Kapitel 6.1 präsentierten Daten verknüpften erstmals den bereits in der Literatur diskutierten Zirkulardichroismus in den Zählraten von Photoionen (PICD) mit dem signalstärkeren differentiellen PICD in der Einfachionisation von Methyloxiran. Dissoziiert das Molekül nach der Ionisation rasch genug, gewährt der Impulsvektor des geladenen Fragments Zugang zu einer Fragmentationsachse. Durch die Auflösung nach einer Molekülachse ist der beobachtete PICD fast eine Größenordnung stärker, als der über alle Raumrichtungen integrierte.
In steigender Komplexität wurde in Kapitel 6.2 eine Fragmentation in vier Teilchen von Molekülen aus einem racemischen Gemisch von CHBrClF untersucht. Über die Auswertung eines Spatproduktes aus den Impulsvektoren konnte für jedes Molekül dessen Händigkeit bestimmt und der vollständig differentielle PICD untersucht werden. Durch das Festhalten einer Fragmentationsachse (analog zu Kapitel 6.1) konnten um einen Faktor vier stärkere PICD-Signale und durch das Auflösen nach der vollständigen Molekülorientierung die Signalstärke des PICD um einen Faktor von etwa 16 in den Bereich einiger Prozente gebracht werden. Leider übersteigt die theoretische Beschreibung dieses Prozesses den aktuellen Stand der Forschung weit. Daher kann nicht ausgeschlossen werden, dass nicht ein Beitrag zur PICD-Signalverstärkung auch aus der Dynamik der sequentiellen vielfachen Ionisation stammt.
Die untersuchte Reaktion in Kapitel 6.3 war der Fünf-Teilchenaufbruch der achiralen Ameisensäure. In der Messung aller ionischen Fragmente konnten analog zu dem vorherigen Kapitel die internen Koordinaten sowie die Orientierung des Moleküls ermittelt werden. Tatsächlich wurde von einer chiralen Fragmentation der achiralen Ameisensäure berichtet. Welches Enantiomer in der Fragmentation beobachtet wird, hängt maßgeblich von der Molekülorientierung relativ zum ionisierenden Laserpuls ab. Diese Erkenntnis könnte zu neuen Ansätzen für Laserkatalysierte enantioselektive Reaktionen führen. Darüber hinaus konnte gezeigt werden, dass die beobachtete Händigkeit des Moleküls nicht nur von seiner Orientierung, sondern auch von der Helizität des ionisierenden Laserpulses abhängt. Dieser differentielle PICD an der Ameisensäure zeigte sich neben einer sehr großen Signalstärke von über 20 % auch als sensitive Probe für die molekulare Struktur.
In Kapitel 7 wurden die Untersuchungen an den 3-dimensionalen Impulsverteilungen der Photoelektronen vorgestellt. Zunächst wird hierzu auf die allgemeine Form des Dichroismus in den Photoelektronen (PECD) im Starkfeldregime eingegangen und die vorherrschenden Symmetrien des Ionisationsregimes herausgearbeitet (Kapitel 7.1). Mit leicht steigender Komplexität konnte eine klare Verbindung zwischen der Asymmetrie in der Elektronenverteilung und dem Schicksal des zurückbleibenden molekularen Ions anhand der Einfachionisation von Methyloxiran herausgearbeitet werden (Kapitel 7.2). Dies hat eine wichtige Auswirkung auf die Nutzbarkeit des PECD im Starkfeldregime als Analysemethode für Chemie und Pharmazie: Der über alle Fragmentationskanäle integrierte PECD ist sensitiv auf die Gewichtung der Fragmente und damit auch auf beispielsweise die maximale Laserintensität. Die Daten legen nahe, dass die Abhängigkeit des PECD von dem Fragmentationskanal auf die unterschiedliche Auswahl von Subensembles molekularer Orientierungen zurückzuführen ist.
Bei Verwendung von elliptisch polarisiertem Licht treten gegenüber der zirkularen Polarisation eine Reihe neuer Effekte auf (Kapitel 7.3). Zunächst zeigt der PECD auch im Starkfeldregime eine nicht lineare Sensitivität auf den Polarisationszustand, welche sich auch als Funktion des Elektronentransversalimpulses und dem Fragmentationskanal ändert. Somit ist die Verwendung von elliptisch polarisiertem Licht bestens für die chirale Erkennung geeignet, wie inzwischen auch in der Literatur bestätigt wurde. Darüber hinaus führt die gebrochene Rotationssymmetrie bei elliptisch polarisiertem Licht zu einer Elektronenimpulsverteilung, welche selbst chiral ist: Der PECD variiert je nach Winkel φ in der Polarisationsebene, wobei die Extrema des PECD nicht mit den Maxima der Zählraten übereinstimmen. Als neue chirale Beobachtungsgröße konnten wir eine enantiosensitive und vorwärts-/rückwärtsasymmetrische Rotation der Zählratenmaxima einführen. Als abgeleitete Größe aus derselben drei-dimensionalen Elektronenverteilung ist diese Beobachtungsgröße jedoch untrennbar verknüpft mit dem ϕ-abhängigen PECD.
Kapitel 8 verknüpfte das (partielle) Wissen um die molekulare Orientierung und den PICD mit den Asymmetrien der Elektronenverteilung für die Messung der fünffach-Ionisation von Ameisensäure (Kapitel 8.1), der vierfach-Ionisation von CHBrClF (Kapitel 8.2) und der Einfachionisation von Methyloxiran (Kapitel 8.3). Im Datensatz der Ameisensäure und dem des CHBrClF zeigte die molekulare Orientierung einen größeren Einfluss auf die Asymmetrie in der Elektronenverteilung als das Enantiomer oder die Helizität des Lichtes. Diese Verknüpfung zwischen Molekülorientierung und Elektronenasymmetrie überträgt die Asymmetrien des PICD auf die Elektronenverteilung. Die Messung an Methyloxiran relativiert diesen Zusammenhang jedoch in dem dieser in dieser Stärke nur bei manchen Fragmentationskanälen auftritt. Offenbar ist die Übertragung der Asymmetrie der differentiellen Ionisationswahrscheinlichkeit nur einer der Mechanismen, welcher zu Elektronasymmetrien im Starkfeldregime führt.
The ruling of the German Federal Constitutional Court and its call for conducting and communicating proportionality assessments regarding monetary policy have been the subject of some controversy. However, it can also be understood as a way to strengthen the de-facto independence of the European Central Bank. The authors shows how a regular proportionality check could be integrated in the ECB’s strategy that is currently undergoing a systematic review. In particular, they propose to include quantitative benchmarks for policy rates and the central bank balance sheet. Deviations from such benchmarks can have benefits in terms of the intended path for inflation while involving costs in terms of risks and side effects that need to be balanced. Seit ihrer Auffindung und Bergung im vergangenen Jahrhundert harren die Bruchstücke dreier Steininschriften aus den Heidelberger Stadtteilen Neuenheim und Handschuhsheim sowie vom Heiligenberg im Depot des Kurpfälzischen Museums Heidelberg ihrer wissenschaftlichen Würdigung. Bei genauerer Betrachtung lassen sie sich trotz ihrer sehr fragmentarischen Erhaltung nahezu zweifelsfrei als Weihinschriften (1. und 2.) beziehungsweise als Grabinschrift (3.) identifizieren, womit sie das Corpus der bereits bekannten epigraphischen Zeugnisse Heidelbergs entsprechend erweitern. Außerdem darf in einer vierten ebenfalls im Depot verwahrten, allerdings bereits publizierten und nur allgemein als Votivstein gedeuteten Inschrift aus Meckesheim / Rhein-Neckar-Kreis (4.) nunmehr so gut wie sicher der Bestandteil einer Jupiter(giganten)säule erkannt werden. Das aus dem Heidelberger Raum bislang dokumentierte Repertoire solcher monumentalen Weihedenkmäler der Römerzeit erfährt damit ebenfalls einen weiteren Zuwachs.
Focused electron beam induced deposition (FEBID) is a direct-write nanofabrication technique able to pattern three-dimensional magnetic nanostructures at resolutions comparable to the characteristic magnetic length scales. FEBID is thus a powerful tool for 3D nanomagnetism which enables unique fundamental studies involving complex 3D geometries, as well as nano-prototyping and specialized applications compatible with low throughputs. In this focused review, we discuss recent developments of this technique for applications in 3D nanomagnetism, namely the substantial progress on FEBID computational methods, and new routes followed to tune the magnetic properties of ferromagnetic FEBID materials. We also review a selection of recent works involving FEBID 3D nanostructures in areas such as scanning probe microscopy sensing, magnetic frustration phenomena, curvilinear magnetism, magnonics and fluxonics, offering a wide perspective of the important role FEBID is likely to have in the coming years in the study of new phenomena involving 3D magnetic nanostructures.
In the course of a growing start-up market and strongly increasing investment volume, investors try to predict the success of a business as precisely as possible in advance. However, when assessing the personality of the founder or founding team, they still rely far too often on their gut feeling, thereby reducing the quality of their decisions. Our study therefore aimed at investigating whether there are any relationships between the founders' personality traits and their performance and thus justifying the need for more targeted and optimized diagnostics in the field of founder personality. With a total of 141 founders, clear correlations between personality traits (conscientiousness, emotional stability) and performance could be demonstrated in the present study. In addition, it became evident that perceived stress is also related to the founders’ personality (emotional stability negative, conscientiousness positive) and in turn has a negative effect on performance. Our findings contribute to raising awareness of the importance of personality as a predictor of founders' performance, improving decision-making, and, in the long run, replacing gut feeling as an inappropriate assessment criterion of investors.
Die UN-Behindertenrechtskonvention bringt unter anderem eine „inklusionsorientierte Lehrer*innenbildung“ hervor, die Lehrkräfte für inklusive Settings kompetent machen soll. Damit das gelingen kann, müssen einerseits für inklusive Settings notwendige Kompetenzen identifiziert und operationalisiert werden, andererseits sind Erkenntnisse darüber notwendig, wie Studierende diese Kompetenzen entwickeln. Erst dann können wirksame Lehr- und Lernformate konzipiert werden. Inzwischen liegen Forschungsergebnisse zu inklusionsbezogenen Aufgaben- und Anforderungsprofilen vor, jedoch über vereinzelte Best-Practice-Beispiele kaum evidenzbasierte Erkenntnisse über Kompetenzentwicklungsprozesse in diesem Bereich oder gar Ableitungen für die Vermittlung solcher Kompetenzen. In diesem Artikel wird für die inklusionsbezogene Teilkompetenz „(multi-)professioneller Kooperationsfähigkeit und -bereitschaft“ ein kompetenzorientiertes Lehr- und Lernformat im Rahmen des Lehramtsstudiums an der Universität Paderborn konzipiert. In Anlehnung an das Constructive-Alignment-Modell werden dafür anhand eines Kompetenzmodells Learning Outcomes definiert, didaktisch-methodische Lernwege zugeordnet und Möglichkeiten einer Evidenzbasierung in Form von Prüfungen und Evaluation diskutiert. Dieser Artikel leistet so einen Beitrag zu einer inklusiven Hochschuldidaktik als Werkzeug, aber auch Voraussetzung und gleichzeitig Ergebnis, einer pädagogischen Fachlichkeit im Spannungsfeld inklusiver Realität.

Recent studies suggest that synaptic lysophosphatidic acids (LPAs) augment glutamate-dependent cortical excitability and sensory information processing in mice and humans via presynaptic LPAR2 activation. Here, we studied the consequences of LPAR2 deletion or antagonism on various aspects of cognition using a set of behavioral and electrophysiological analyses. Hippocampal neuronal network activity was decreased in middle-aged LPAR2−/− mice, whereas hippocampal long-term potentiation (LTP) was increased suggesting cognitive advantages of LPAR2−/− mice. In line with the lower excitability, RNAseq studies revealed reduced transcription of neuronal activity markers in the dentate gyrus of the hippocampus in naïve LPAR2−/− mice, including ARC, FOS, FOSB, NR4A, NPAS4 and EGR2. LPAR2−/− mice behaved similarly to wild-type controls in maze tests of spatial or social learning and memory but showed faster and accurate responses in a 5-choice serial reaction touchscreen task requiring high attention and fast spatial discrimination. In IntelliCage learning experiments, LPAR2−/− were less active during daytime but normally active at night, and showed higher accuracy and attention to LED cues during active times. Overall, they maintained equal or superior licking success with fewer trials. Pharmacological block of the LPAR2 receptor recapitulated the LPAR2−/− phenotype, which was characterized by economic corner usage, stronger daytime resting behavior and higher proportions of correct trials. We conclude that LPAR2 stabilizes neuronal network excitability upon aging and allows for more efficient use of resting periods, better memory consolidation and better performance in tasks requiring high selective attention. Therapeutic LPAR2 antagonism may alleviate aging-associated cognitive dysfunctions.
Context information supports serial dependence of multiple visual objects across memory episodes

Serial dependence is thought to promote perceptual stability by compensating for small changes of an object’s appearance across memory episodes. So far, it has been studied in situations that comprised only a single object. The question of how we selectively create temporal stability of several objects remains unsolved. In a memory task, objects can be differentiated by their to-be-memorized feature (content) as well as accompanying discriminative features (context). We test whether congruent context features, in addition to content similarity, support serial dependence. In four experiments, we observe a stronger serial dependence between objects that share the same context features across trials. Apparently, the binding of content and context features is not erased but rather carried over to the subsequent memory episode. As this reflects temporal dependencies in natural settings, our findings reveal a mechanism that integrates corresponding content and context features to support stable representations of individualized objects over time.
The yeast fatty acid synthase (FAS) is a barrel-shaped 2.6 MDa complex. Upon barrel-formation, two multidomain subunits, each more than 200 kDa large, intertwine to form a heterododecameric complex that buries 170,000 Å2 of protein surface. In spite of the rich knowledge about yeast FAS in structure and function, its assembly remained elusive until recently, when co-translational interaction of the β-subunit with the nascent α-subunit was found to initiate assembly. Here, we characterize the co-translational assembly of yeast FAS at a molecular level. We show that the co-translationally formed interface is sensitive to subtle perturbations, so that the exchange of two amino acids located in the emerging interface can prevent assembly. On the other hand, assembly can also be initiated via the co-translational interaction of the subunits at other sites, which implies that this process is not strictly site or sequence specific. We further highlight additional steps in the biogenesis of yeast FAS, as the formation of a dimeric subunit that orchestrates complex formation and acts as platform for post-translational phosphopantetheinylation. The presented data supports the understanding of the recently discovered prevalence of eukaryotic complexes for co-translational assembly, and is valuable for further harnessing FAS in the biotechnological production of aliphatic compounds.
The transition from local to global patterns governs the differentiation of mouse blastocysts

During mammalian blastocyst development, inner cell mass (ICM) cells differentiate into epiblast (Epi) or primitive endoderm (PrE). These two fates are characterized by the expression of the transcription factors NANOG and GATA6, respectively. Here, we investigate the spatio-temporal distribution of NANOG and GATA6 expressing cells in the ICM of the mouse blastocysts with quantitative three-dimensional single cell-based neighbourhood analyses. We define the cell neighbourhood by local features, which include the expression levels of both fate markers expressed in each cell and its neighbours, and the number of neighbouring cells. We further include the position of a cell relative to the centre of the ICM as a global positional feature. Our analyses reveal a local three-dimensional pattern that is already present in early blastocysts: 1) Cells expressing the highest NANOG levels are surrounded by approximately nine neighbours, while 2) cells expressing GATA6 cluster according to their GATA6 levels. This local pattern evolves into a global pattern in the ICM that starts to emerge in mid blastocysts. We show that FGF/MAPK signalling is involved in the three-dimensional distribution of the cells and, using a mutant background, we further show that the GATA6 neighbourhood is regulated by NANOG. Our quantitative study suggests that the three-dimensional cell neighbourhood plays a role in Epi and PrE precursor specification. Our results highlight the importance of analysing the three-dimensional cell neighbourhood while investigating cell fate decisions during early mouse embryonic development.
Phytochrome photoreceptors operate via photoisomerization of a bound bilin chromophore. Their typical architecture consists of GAF, PAS and PHY domains. Knotless phytochromes lack the PAS domain, while retaining photoconversion abilities, with some being able to photoconvert with just the GAF domain. Therefore, we investigated the ultrafast photoisomerization of the Pr state of a knotless phytochrome to reveal the effect of the PHY domain and its “tongue” region on the transduction of the light signal. We show that the PHY domain does not affect the initial conformational dynamics of the chromophore. However, it significantly accelerates the consecutively induced reorganizational dynamics of the protein, necessary for the progression of the photoisomerization. Consequently, the PHY domain keeps the bilin and its binding pocket in a more reactive conformation, which decreases the extent of protein reorganization required for the chromophore isomerization. Thereby, less energy is lost along nonproductive reaction pathways, resulting in increased efficiency.
The impact of the appropriate and inappropriately applied statistical metrics to verify the State of Control of pharmaceutical manufacturing has been reviewed from an auditor’s perspective. Good and bad statistical practices have been presented in an attempt for manufactures to appreciate the risks of using these metrics. Conclusions concerning (1) control charts to be used instead of line/run charts for trend analysis, (2) Ppk as the preferred capability index (but still with an ambition to get processes into statistical control), (3) show process capability indices along with their respective control charts (4) determine which Manufacturing State of Control the product/process lies in (5) an effective Control Strategy can only be implemented if the Manufacturing State of Control is understood, (6) when presenting data consider what is truly representative of the product/process and not the average (7) Management should align with ICH Q10 more effectively to provide statistical resources for their personnel.
Effective connectivity (EC) is able to explore causal effects between brain areas and can depict mechanisms that underlie repair and adaptation in chronic brain diseases. Thus, the application of EC techniques in multiple sclerosis (MS) has the potential to determine directionality of neuronal interactions and may provide an imaging biomarker for disease progression. Here, serial longitudinal structural and resting-state fMRI was performed at 12-week intervals over one year in twelve MS patients. Twelve healthy subjects served as controls (HC). Two approaches for EC quantification were used: Causal Bayesian Network (CBN) and Time-resolved Partial Directed Coherence (TPDC). The EC strength was correlated with the Expanded Disability Status Scale (EDSS) and Fatigue Scale for Motor and Cognitive functions (FSMC). Our findings demonstrated a longitudinal increase in EC between specific brain regions, detected in both the CBN and TPDC analysis in MS patients. In particular, EC from the deep grey matter, frontal, prefrontal and temporal regions showed a continuous increase over the study period. No longitudinal changes in EC were attested in HC during the study. Furthermore, we observed an association between clinical performance and EC strength. In particular, the EC increase in fronto-cerebellar connections showed an inverse correlation with the EDSS and FSMC. Our data depict continuous functional reorganization between specific brain regions indicated by increasing EC over time in MS, which is not detectable in HC. In particular, fronto-cerebellar connections, which were closely related to clinical performance, may provide a marker of brain plasticity and functional reserve in MS.
Psychotherapists in mental health institutions as a professional group are part of the medical system, and from this perspective, as representing an occupation that serves the public health interests, as well as those of the individual seeking help. Despite the different existing therapeutic approaches and diverse forms of therapy deriving from these approaches critical theories, however, consider psychotherapy as a profession with a specific jurisdictional claim and own highly specific interests. In contrast to most of the recent discussion around therapy culture, in this article, I argue that sociology and social theory could benefit from an understanding of psychotherapy as a profession with a separate logic and claim for jurisdiction for mental health. Moreover, I present some general trends showing that, regarding psychotherapy, we face a concurrence of a professionalisation, and simultaneously, an already ongoing deprofessionalisation. To develop my argument, I first discuss the perspectives of sociology of the psychotherapy professions. Second, I present the potential lack of professionalism in four dimensions. Third, I discuss possible tendencies of deprofessionalisation. Finally, I conclude by pointing out the importance of theorising the psychotherapy professions for medical sociology.
Objectives: The ongoing coronavirus pandemic is challenging, especially in severely affected patients who require intubation and sedation. Although the potential benefits of sedation with volatile anesthetics in coronavirus disease 2019 patients are currently being discussed, the use of isoflurane in patients with coronavirus disease 2019–induced acute respiratory distress syndrome has not yet been reported. Design: We performed a retrospective analysis of critically ill patients with hypoxemic respiratory failure requiring mechanical ventilation. Setting: The study was conducted with patients admitted between April 4 and May 15, 2020 to our ICU. Patients: We included five patients who were previously diagnosed with severe acute respiratory syndrome coronavirus 2 infection. Intervention: Even with high doses of several IV sedatives, the targeted level of sedation could not be achieved. Therefore, the sedation regimen was switched to inhalational isoflurane. Clinical data were recorded using a patient data management system. We recorded demographical data, laboratory results, ventilation variables, sedative dosages, sedation level, prone positioning, duration of volatile sedation and outcomes. Measurements & Main Results: Mean age (four men, one women) was 53.0 (± 12.7) years. The mean duration of isoflurane sedation was 103.2 (± 66.2) hours. Our data demonstrate a substantial improvement in the oxygenation ratio when using isoflurane sedation. Deep sedation as assessed by the Richmond Agitation and Sedation Scale was rapidly and closely controlled in all patients, and the subsequent discontinuation of IV sedation was possible within the first 30 minutes. No adverse events were detected. Conclusions: Our findings demonstrate the feasibility of isoflurane sedation in five patients suffering from severe coronavirus disease 2019 infection. Volatile isoflurane was able to achieve the required deep sedation and reduced the need for IV sedation.
Proper speech production requires auditory speech feedback control. Models of speech production associate this function with the right cerebral hemisphere while the left hemisphere is proposed to host speech motor programs. However, previous studies have investigated only spectral perturbations of the auditory speech feedback. Since auditory perception is known to be lateralized, with right-lateralized analysis of spectral features and left-lateralized processing of temporal features, it is unclear whether the observed right-lateralization of auditory speech feedback processing reflects a preference for speech feedback control or for spectral processing in general. Here we use a behavioral speech adaptation experiment with dichotically presented altered auditory feedback and an analogous fMRI experiment with binaurally presented altered feedback to confirm a right hemisphere preference for spectral feedback control and to reveal a left hemisphere preference for temporal feedback control during speaking. These results indicate that auditory feedback control involves both hemispheres with differential contributions along the spectro-temporal axis.
This article investigates the L1 acquisition of different types of direct objects in European Portuguese (EP). Previous research has revealed that although children have early syntactic and pragmatic knowledge of objects across languages, the adequate use of pronouns and null objects is protracted in the acquisition of EP (Costa et al. 2012). The present study shows that children acquiring the distribution of direct objects are aware of universal pragmatic hierarchies but struggle with the interpretation and feature bundles of null objects. Assuming that arguments are linked to left-peripheral C/edge linkers (Sigurðsson 2011), we argue that children need more time to discover the adult-like feature composition of null objects in EP because they involve phi-silent features. Relative accessibility (Ariel 1991) is universal and available early, whereas the absolute accessibility of null objects, i.e. their feature content, is acquired relatively late.
Background: Marked sex differences in autism prevalence accentuate the need to understand the role of biological sex-related factors in autism. Efforts to unravel sex differences in the brain organization of autism have, however, been challenged by the limited availability of female data.
Methods: We addressed this gap by using a large sample of males and females with autism and neurotypical (NT) control individuals (ABIDE; Autism: 362 males, 82 females; NT: 409 males, 166 females; 7-18 years). Discovery analyses examined main effects of diagnosis, sex and their interaction across five resting-state fMRI (R-fMRI) metrics (voxel-level Z > 3.1, cluster-level P < 0.01, gaussian random field corrected). Secondary analyses assessed the robustness of the results to different pre-processing approaches and their replicability in two independent samples: the EU-AIMS Longitudinal European Autism Project (LEAP) and the Gender Explorations of Neurogenetics and Development to Advance Autism Research (GENDAAR).
Results: Discovery analyses in ABIDE revealed significant main effects across the intrinsic functional connectivity (iFC) of the posterior cingulate cortex, regional homogeneity and voxel-mirrored homotopic connectivity (VMHC) in several cortical regions, largely converging in the default network midline. Sex-by-diagnosis interactions were confined to the dorsolateral occipital cortex, with reduced VMHC in females with autism. All findings were robust to different pre-processing steps. Replicability in independent samples varied by R-fMRI measures and effects with the targeted sex-by-diagnosis interaction being replicated in the larger of the two replication samples – EU-AIMS LEAP.
Limitations: Given the lack of a priori harmonization among the discovery and replication datasets available to date, sample-related variation remained and may have affected replicability.
Conclusions: Atypical cross-hemispheric interactions are neurobiologically relevant to autism. They likely result from the combination of sex-dependent and sex-independent factors with a differential effect across functional cortical networks. Systematic assessments of the factors contributing to replicability are needed and necessitate coordinated large-scale data collection across studies.

To make progress we need to both identify and validate neural markers that help to parse heterogeneity to tailor therapies to specific neurobiological profiles. Atypical hemispheric lateralization is a stable feature across studies in autism, but its potential as a neural stratification marker has not been widely examined. Methods: In order to dissect heterogeneity in lateralization in autism, we used the large EU-AIMS (European Autism Interventions—A Multicentre Study for Developing New Medications) Longitudinal European Autism Project dataset comprising 352 individuals with autism and 233 neurotypical control subjects as well as a replication dataset from ABIDE (Autism Brain Imaging Data Exchange) (513 individuals with autism, 691 neurotypical subjects) using a promising approach that moves beyond mean group comparisons. We derived gray matter voxelwise laterality values for each subject and modeled individual deviations from the normative pattern of brain laterality across age using normative modeling. Results: Individuals with autism had highly individualized patterns of both extreme right- and leftward deviations, particularly in language, motor, and visuospatial regions, associated with symptom severity. Language delay explained most variance in extreme rightward patterns, whereas core autism symptom severity explained most variance in extreme leftward patterns. Follow-up analyses showed that a stepwise pattern emerged, with individuals with autism with language delay showing more pronounced rightward deviations than individuals with autism without language delay. Conclusions: Our analyses corroborate the need for novel (dimensional) approaches to delineate the heterogeneous neuroanatomy in autism and indicate that atypical lateralization may constitute a neurophenotype for clinically meaningful stratification in autism.

This article elucidates the spatial order that underpins the politics of the Anthropocene – the ecological nomos of the earth – and criticizes its imperial origins and legacies. It provides a critical reading of Carl Schmitt’s spatial thought to not only illuminate the spatio-political ontology but also the violence and usurpations that characterize the Anthropocene condition. The article first shows how with the emergence of the ecological nomos seemingly ‘natural’ spaces like the biosphere and the atmosphere became politically charged. This challenges the modernist separation between natural facts and political norms. It then underlines the imperial origins of this nomos by introducing the concept of air-appropriation understood as the colonization of atmospheric space by CO2 emissions. Instead of assuming that the ecological nomos represents a transition from a colonial to an ecological and cosmopolitan world order, focusing on air-appropriation highlights forms of ecological imperialism that go along with the new nomos. Accordingly, the article calls for a just redistribution of ecospace that takes into account the imperial legacies and ongoing effects of air-appropriation.
The Izu–Bonin–Mariana volcanic arc is situated at a convergent plate margin where subduction initiation triggered the formation of MORB-like forearc basalts as a result of decompression melting and near-trench spreading. International Ocean Discovery Program (IODP) Expedition 352 recovered samples within the forearc basalt stratigraphy that contained unusual macroscopic globular textures hosted in andesitic glass (Unit 6, Hole 1440B). It is unclear how these andesites, which are unique in a stratigraphic sequence dominated by forearc basalts, and the globular textures therein may have formed. Here, we present detailed textural evidence, major and trace element analysis, as well as B and Sr isotope compositions, to investigate the genesis of these globular andesites. Samples consist of K2O-rich basaltic globules set in a glassy groundmass of andesitic composition. Between these two textural domains a likely hydrated interface of devitrified glass occurs, which, based on textural evidence, seems to be genetically linked to the formation of the globules. The andesitic groundmass is Cl rich (ca. 3000 µg/g ), whereas globules and the interface are Cl poor (ca. 300 µg/g ). Concentrations of fluid-mobile trace elements also appear to be fractionated in that globules and show enrichments in B, K, Rb, Cs, and Tl, but not in Ba and W relative to the andesitic groundmass, whereas the interface shows depletions in the latter, but is enriched in the former. Interestingly, globules and andesitic groundmass have identical Sr isotopic composition within analytical uncertainty ( 87Sr∕86Sr of 0.70580 ± 10 ), indicating that they likely formed from the same source. However, globules show high δ11 B (ca. + 7 ‰ ), whereas their host andesites are isotopically lighter (ca. – 1 ‰ ), potentially indicating that whatever process led to their formation either introduced heavier B isotopes to the globules, or induced stable isotope fractionation of B between globules and their groundmass. Based on the bulk of the textural information and geochemical data obtained from these samples, we conclude that these andesites likely formed as a result of the assimilation of shallowly altered oceanic crust (AOC) during forearc basaltic magmatism. Assimilation likely introduced radiogenic Sr, as well as heavier B isotopes to comparatively unradiogenic and low δ11B forearc basalt parental magmas (average 87Sr∕86Sr of 0.703284). Moreover, the globular textures are consistent with their formation being the result of fluid-melt immiscibility that was potentially induced by the rapid release of water from assimilated AOC whose escape likely formed the interface. If the globular textures present in these samples are indeed the result of fluid-melt immiscibility, then this process led to significant trace element and stable isotope fractionation. The textures and chemical compositions of the globules highlight the need for future experimental studies aimed at investigating the exsolution process with respect to potential trace element and isotopic fractionation in arc magmas that have perhaps not been previously considered.
Due to anticipated postoperative neuropsychological sequelae, patients with gliomas infiltrating the corpus callosum rarely undergo tumor resection and mostly present in a poor neurological state. We aimed at investigating the benefit of glioma resection in the corpus callosum, hypothesizing neuropsychological deficits were mainly caused by tumor presence. Between 01/2017 and 1/2020, 21 patients who underwent glioma resection in the corpus callosum were prospectively enrolled into this study. Neuropsychological function was assessed preoperatively, before discharge and after 6 months. Gross total tumor resection was possible in 15 patients, and in 6 patients subtotal tumor resection with a tumor reduction of 97.7% could be achieved. During a median observation time of 12.6 months 9 patients died from glioblastoma after a median of 17 months. Preoperatively, all cognitive domains were affected in up to two thirds of patients, who presented a median KPS of 100% (range 60–100%). After surgery, the proportion of impaired patients increased in all neurocognitive domains. Most interestingly, after 6 months, significantly fewer patients showed impairments in attention, executive functioning, memory and depression, which are domains considered crucial for everyday functionality. Thus, the results of our study strongly support our hypothesis that in patients with gliomas infiltrating the corpus callosum the benefit of tumor resection might outweigh morbidity.
