Ischemic injuries of the cardiovascular system are still the leading cause of death worldwide. They are often accompanied by loss of cardiomyocytes (CM) and their replacement by non-functional heart tissue. Cardiac fibroblasts (CF) play a major role in the recovery after ischemic injury and in the scar formation. In the last few years researchers were able to reprogram fibroblasts into CM in vitro and in murine models of myocardial infarction using various protocols including a cocktail of microRNAs (miRs). These miRs can target hundreds of messenger RNAs and inhibit their translation into proteins, potentially regulating multiple cellular signaling pathways. Because of this, there has been a rising interest in the use of miRs for therapeutic purposes. However, as different miRs have different effects in different cells, there is the danger of causing serious side effects. These could be alleviated by enacting a cell-specific transport of miRs, for example by using aptamers. Aptamers are usually short strands of DNA or RNA, which can fold into a specific three-dimensional confirmation which allows them to bind specifically to target molecules. Aptamers are commonly selected from a large library for their ability to bind to target molecules using a procedure called SELEX. Aptamers have already been used to transport miRs into cancer cells.
In this thesis, we first established the transport of miRs into cells of the cardiovascular system using aptamers. MiR-126 is an important part of the signaling in endothelial cells (EC), protects from atherosclerosis and supports angiogenesis, which is why we chose it as a candidate to transport into the vasculature. We first tested two aptamers for their ability to internalize into EC and fibroblasts. Both the aptamer for the ubiquitously expressed transferrin receptor (TRA) and a general internalizing RNA motif, but not a control construct, could internalize efficiently into all cell types tested. We then designed three chimeras (Ch) using different strategies to connect TRA to miR-126. While all chimeras could internalize efficiently, only Ch3, which connects TRA to Pre-miR-126 using a sticky bridge structure, had functional effects in EC. Ch3 reduced the protein expression of VCAM-1 in EC and increased the VEGF induced sprouting of EC in a spheroid-sprouting assay. Treatment of breast cancer cells with Ch3 emulated the effects of treatment with classical miR-126-3p and miR-126-5p mimics. In the SK-BR3 cell line Ch3 and miR-126-3p reduce the viability of the cells while they reduce recruitment of EC by the MCF7 cell line. miR-126-5p had no apparent effect in the SK-BR3 line, but increased viability of MCF7 cells, as did Ch3. This implies that Ch3 can be processed to both functional miR-126-3p and miR-126-5p in treated cells.
We were unable to achieve a reprogramming of adult murine cardiac fibroblasts into cells resembling CM using the cocktail of 4 miRs. This indicates that the miR-mediated transdifferentiation is only possible in neonatal fibroblasts. The effects in mice after an AMI might possibly be caused by an enhanced plasticity of fibroblasts in and close to the infarcted area.
We also screened to find aptamers specifically binding to cells of the cardiovascular system. We used two oligonucleotide libraries in a cell-SELEX to select candidates which bind to CF, but not EC. We observed that only the library which contains two randomized regions of 26 bases showed an enrichment of species binding to fibroblasts. We then sequenced rounds 5-7 of the SELEX and analyzed the data bioinfomatically to select 10 candidate aptamers. All candidates showed a strong binding not only to CF, but also EC. This indicates that the selection pressure against species binding to EC was not high enough and would have to be increased to find true CF-aptamers. Four promising candidates were also analyzed for their potential to be internalized and we surprisingly found that all of them were internalized by EC and CF more efficiently than TRA. The similar behavior of the candidates implies that they possibly share a ligand, which is expressed both by EC and CF, but more prominently by the latter.
This work demonstrates the possibility of using aptamers to transport miRs into cells of the cardiovascular system. It also shows that it is possible to select aptamers for non-cancerous mammalian cells, which has not been done before. It is reasonable to assume that a refinement of the cell-SELEX will allow selection of cell-specific aptamers. Due to the failure of reprogramming of adult fibroblasts into induced cardiomyocytes we were unable to test whether a miR-mediated reprogramming might be inducible using aptamer transported-miRs. Ultimately, aptamer mediated transport of miRs is a feasible and promising therapeutic option for the treatment of cardiovascular diseases and other disorders like cancer.
The development of the atrioventricular (AV) canal and the cardiac valves is tightly linked and a critically regulated process. Anomalies in components of the involved pathways can lead to congenital valve malformations, a leading cause of morbidity and mortality in neonates. Myocardial Bmp as well as endocardial Notch and Wnt signaling have been identified as critical factors for the induction of EMT during the formation of the endocardial cushions and cardiac valves. Of these, canonical Wnt signaling positively regulates endocardial proliferation and EMT but negatively regulates endocardial differentiation. Further, elevated Wnt signaling leads to the ectopic expression of myocardial Bmp ligands suggesting a high level of integration of the involved pathways and crosstalk amongst the different cardiac tissues.
Here we have identified a novel role for Id4 as a mediator between Bmp and Wnt signaling. Id4 belongs to the Id family of proteins and is known to be involved in bone and nervous system development. We found that in zebrafish, id4 is expressed in the endocardium of the AV canal at embryonic stages and throughout the atrial chamber in addition to AV canal, in adults. Using transcription activator-like effector nucleases (TALENs) we established an id4 mutant allele. Our analysis shows that id4 mutant larvae are susceptible to retrograde blood flow, and show aberrant expression of developmental valvular markers. These include expanded expression domains of markers like bmp4, cspg2a and Alcam. In contrast, valve maturation as assessed by the expression of spp1 is considerably reduced in id4 mutants. Using conditional transgenic systems, along with elegant in vivo imaging of transgenic reporter lines, we further found that id4 is a transcriptional target of Bmp signaling, and it is capable of dose dependently restricting Wnt signaling in the endocardium of the Atrioventricular Canal.
Taken together, our data identifies Id4 as a novel player in Atrioventricular Canal and valve development. We show that Id4 function is important in valve development acting downstream of Bmp signaling by restricting endocardial Wnt to allow valve maturation
Die vorliegende Studie vermittelt einen epidemiologischen Überblick über das mit Haut- und Nagelläsionen assoziierte Pilzspektrum im Westen Panamas. Hierzu wurden Proben von vermutlich durch Pilzinfektionen verursachten Haut- sowie Nagelläsionen gesammelt und zum Anlegen von Kulturen verwendet. Die isolierten Pilze wurden basierend auf dem D-H-S System (Rieth), anhand morphologischer Merkmale, rDNA Sequenzdaten sowie phylogenetischen Analysen klassifiziert und mit Hilfe von Literaturdaten sowie physiologischen Eigenschaften als saprotrophe, opportunistische oder pathogene Organismen beurteilt. In Panama wurden 52 Proben von 51 Personen gesammelt, wobei das Material von 42 Haut- und Nagelläsionen der Füße, vier Läsionen der Fingernägel, zwei Chromomykosen, einer Tinea nigra und drei sonstigen Hautläsionen stammt. Bei 75 Prozent (n = 39) der Proben konnten Pilze kultiviert und insgesamt 201 Pilzstämme isoliert und subkultiviert werden. Hiervon wurden 50 Isolate (24,9 %) als Dermatophyten, 24 Stämme (11,9 %) als Hefen und 127 Isolate (63,2 %) als Schimmelpilze klassifiziert. Bei 19 Probanden (48,7 %) konnten Dermatophyten isoliert werden, wobei aus dem Probenmaterial von 12 Personen (63,2 %) ebenfalls andere Pilzarten nachgewiesen wurden. Von zwei Läsionen (5,1 %) wurden nur Hefen isoliert, wobei einmal eine Schwarze Hefe kultiviert wurde. In dem Material acht weiterer Proben (20,5 %) wurden Schimmelpilze und Hefestämme nachgewiesen und bei zehn Probanden (25,6 %) konnten aus dem Probenmaterial nur Schimmelpilze kultiviert werden. 172 Isolate wurden taxonomisch klassifiziert und 44 Arten aus 25 Gattungen, 17 Familien, 15 Ordnungen, sechs Klassen sowie den Abteilungen Ascomycota oder Basidiomycota zugeordnet. Die Ascomyceten stellen mit 164 Stämmen 40 verschiedener Arten aus 23 Gattungen, 15 Familien, 11 Ordnungen und vier Klassen die am häufigsten isolierte und vielfältigste Gruppe dar, während die Basidiomycota nur mit acht Isolaten vier verschiedener Arten zwei unterschiedlicher Gattungen, Familien, Ordnungen und Klassen nachgewiesen wurden. Im Rahmen dieser Arbeit wurden in Panama die anthropophilen Dermatophyten Trichophyton rubrum und T. interdigitale dokumentiert, wobei T. rubrum die am häufigsten isolierte Art darstellt. Kultivierte Hefen waren Candida albicans, C. duobushaemulonii, C. tropicalis, Hortaea werneckii, Sporobolomyces sp., Trichosporon asahii, T. japonicum und T. montevideense. Die Schimmelpilze stellen die größte und ökologisch diverseste Organismengruppe der kultivierten Pilze dar. So wurden von den untersuchten Läsionen sowohl humanpathogene Erreger, als auch opportunistische Arten und rein saprotrophe Pilze sowie mehrere Vertreter wahrscheinlich bisher nicht wissenschaftlich beschriebener Arten bzw. Gattungen nachgewiesen. Aus dem Probenmaterial wurden die Pilze Acremonium collariferum, Aspergillus awamori, A. clavatus, A. flavus, A. giganteus, A. heteromorphus, A. niger, A. ochraceus, A. sclerotiorum, A. versicolor, Chaetomium globosum, Chrysosporium tuberculatum, Cladosporium sphaerospermum, C. tenuissimum, Curvularia geniculata, C. lunata, Fonsecaea pedrosoi, Fusarium oxysporum, F. solani, Lophotrichus bartlettii, Microascus cinereus, Neoscytalidium dimidiatum, Penicillium commune, Scolecobasidium sp., Scopulariopsis carbonaria, S. croci, Verticillium cf. epiphytum und Wardomycopsis litoralis isoliert. Zudem wurden vier Isolate von zwei vermutlich neuen Arten der Gattung Acremonium (Bionectriaceae, Hypocreales), zwei Stämme mit einer genetischen Affinität zu der Gattung Cryptendoxyla (Cephalothecaceae, Sordariales) und jeweils ein mit den Gattungen Fusicladium (Venturiaceae, Venturiales), Knufia (Trichomeriaceae, Chaetothyriales) bzw. Rhexothecium (Eremomycetaceae, Dothideomycetidae) assoziierter Stamm kultiviert. Im Rahmen dieser Studie wurden A. giganteus, C. tenuissimum, L. bartlettii, S. carbonaria, S. croci, V. epiphytum und W. litoralis erstmalig von Mykosen des Menschen dokumentiert und die in der Literatur als Verursacher sowie Besiedler von Haut- und Nagelläsionen beschriebenen Organismen A. clavatus, A. flavus, A. niger, A. ochraceus, C. tropicalis, C. globosum, C. sphaerospermum, C. lunata, F. oxysporum, M. cinereus, P. commune, T. asahii, T. japonicum und T. montevideense wurden das erste Mal in klinischem Probenmaterial aus Panama nachgewiesen. Die Arten A. awamori, A. heteromorphus, C. globosum, C. tenuissimum, L. bartlettii, M. cinereus, P. commune, S. croci, T. asahii, T. japonicum, T. montevideense, V. epiphytum, W. litoralis und die Gattung Scolecobasidium wurden zudem erstmalig für Panama dokumentiert. Die Isolation von W. litoralis ist ebenfalls der erste Nachweis dieses Pilzes außerhalb von Spanien und auf dem amerikanischen Kontinent. Die große Anzahl im Rahmen dieser Arbeit beschriebener, bisher für die Wissenschaft unbekannter bzw. nicht in Panama dokumentierter Pilzarten lässt auf eine große mykologische Biodiversität in Panama schließen und zeigt den Bedarf weiterer Forschung.
Panama, a small country between the major continents of North and South America, is one of the lesser studied regions in Central America, but is recognized for its mega-biodiversity. This is particularly true for Eastern Panama, which I am considering as the easternmost portion of the country, covering the area from the Chepo, which is also the beginning of the San Blas mountain range, towards east, up to the Darien Mountain range on the border with its neighboring country Colombia. In the lowland region I visited two physiographic areas: the Isthmian-Atlantic Moist Forests (IAMF) and the Chocó-Darién Moist Forests (CDMF). In the IAMF I worked at the localities of Río Mono, Wacuco, La Moneda, Arretí, Metetí, Filo del Tallo, and Laguna de Matusagaratí. In the CDMF I visited the localities of Cruce de Mono, Cana, Garachiné, Sambú, and Pavarandó. And I have worked in the highlands of Darién (DM), Majé (MM), Jingurudó-Sapo (JSM), Pirre (PM) and San Blas (SSM) in the highlands.
Before my research, 138 reptile and 104 amphibian species had been reported for EP. From 2008 to 2013, I collected specimens to evaluate the diversity of amphibians and reptiles for this region. I applied an integrative approach to evaluate the taxonomy, diversity, biogeography, and conservation of the herpetofauna of EP. I included analyses of morphometrics, molecular genetics (e.g. barcoding), biogeography, bioacoustics (in anurans), hemipenial morphology (in squamates), and ecology. This is the first regional evaluation of the biodiversity in EP applying integrative taxonomy. Aside from morphological and bioacoustic data, my work is based on the barcoding of 608 specimens, from which I obtained 16S mtDNA for 486 specimens and COI mtDNA for 455. In total I have got sequences for 69.2 %of the amphibian and 48.6 % of the reptile species present in EP. For the morphological analyses, I compared 1597 specimens, including my samples complemented by specimens obtained from various museums. The bioacoustic data were obtained from the analysis of 1504 calls of 27 species of frogs. Based on specimens collected in EP and according to external morphology, I could identify 65 species of amphibians and 72 reptiles, but after applying an integrative approach these numbers increased to 79 amphibians and 88 reptiles described species within my collected specimens. Additionally, I uncovered 33 taxonomic units that could not be assigned to any described species until now, 22 of them represent confirmed candidate species (CCS), and 11 were classified as Unconfirmed candidate species (UCS). Thus, increasing the known species of amphibian by 19.4 % and of reptiles by 4.8 %. Currently, there are 145 reptiles and 129 amphibians known to occur in EP. Based on my results, I have initiated several projects to solve taxonomic uncertanties, including the species of the genera Bolitoglossa, Diasporus, Dactyloa, Ecnomiohyla, Lepidoblepharis, and the taxonomic status of the species Pristimantis caryophyllaceus and Norops tropidogaster.
Out of the 22 CCS I found, I described nine species new to science with type locality in EP, six amphibians and four reptiles. Among these is a new species of Bolitoglossa described from Cerro Chucantí, Cordillera de Majé, Provincia de Darién, Panama. Additionally, I include comments on the other species of congeneric salamanders known to occur in the region. Among the tink frogs, only Diasporus quidditus was known to occur in EP. During my field work I collected six additional species of this genus, four of which are new to science, plus two species new for this region.
I also described one new species of Dactyloa (giant anole lizards) related to the former D. chocorum. I synonymized D. chocorum with D. purpurescens, and included information about the other species of the group from EP. The new species of Dactyloa resembles D. ibanezi, D. limon, and D. purpurescens in external morphology but differs from these species in dewlap coloration, dorsal color pattern, morphometrics, and scalation. I discovered one species of the genus Ecnomiohyla, which exhibits significant genetic distances (16S mtDNA gene) and morphological differences to all known Ecnomiohyla species. Along with the description of the new Ecnomiohyla species, I provide detailed comparisons of morphological and molecular characters of almost all members of the genus in Lower Central America, as well as an identification key for the entire genus. Two new species of the genus Lepidoblepharis from EP were described. In the corresponding work, I include an analysis of Lepidoblepharis spp. in the region, including phylogeography and taxonomy. One of the new species, Lepidoblepharis emberawoundule, can be differentiated from most species in the genus by its small size and its low number of lamellae under the fourth toe and finger. The other species described from EP, Lepidoblepharis rufigularis, can be differentiated from all species in the genus by its small size and the reddish throat in males.
Recently, two of the most common types of bone cancers in children and young adults have been proven to exhibit vulnerability to poly(ADP)-ribose polymerase, (PARP) inhibitors (e.g. olaparib, talazoparib). Ewing’s sarcoma (ES) are reported to harbor a fusion gene EWS-FLI1 (85%), inducing tumorigenesis. Additional, as the fusion gene acts as aberrant transcription factor, it similarly induces elevated PARP expression levels sensitizing ES to PARP inhibition. Second, by an exome sequencing approach in a set of primary osteosarcomas (OS) we identified mutation signatures being reminiscent of BRCA deficiency. Therefore, the sensitivity of a panel of OS cell lines to either talazoparib single treatment or in combination with several chemotherapeutic drugs was investigated.
To screen ES tumor cell lines against PARP inhibitors we applied four different PARP inhibitors (talazoparib, olaparib, niraparib and veliparib) that are frequently being used for clinical studies. We combined those PARP inhibitors with a set of chemotherapeutics (temozolomide (TMZ), SN-38, etoposide, ifosfamide, doxorubicin, vincristine and actinomycin D) that are part of the first-line therapy of ES patients. Here, we demonstrate how PARP inhibitors synergize with TMZ or SN-38 to induce apoptosis, whereas the combination of PARP inhibitors with the other drugs are not favorable. By investigation of key checkpoints in the molecular mechanisms of cell death, the pivotal role of the mitochondrial pathway of apoptosis mediating the synergy between olaparib and TMZ was revealed.
Employing talazoparib monotherapy in combination with or without several chemotherapeutic drugs (TMZ, SN-38, cisplatin, doxorubicin, methotrexate and etoposide/carboplatin), the correlation between homologous recombination (HR) repair deficiency (BRCAness) and the response to talazoparib as prototypical PARP inhibitor was validated in different OS cell lines. By calculation of combination indices (CI) and fraction affected (Fa) values, we identified TMZ as the most potent chemotherapeutic drug in combination with talazoparib inducing the mitochondrial apoptotic pathway in OS.
In our studies of two independent tumor entities with contrary genetic background we identified the combination of PARP inhibitor and TMZ as being most effective. Our studies point out that after TMZ induced DNA methylation and concomitant PARP trapping, DNA damage-imposed checkpoint kinase activation consequently induces G2-cell cycle arrest. Subsequent, PARP inhibitor/TMZ causes MCL-1 degradation, followed by activation of BAK and BAX, succeeding in loss of mitochondrial outer membrane potential (LMMP) and activation of downstream effector-caspases in mitochondrial apoptosis. Our findings emphasize the importance of PARP inhibition in order to chemosensitize ES, which express high PARP levels, or OS that bear features of BRCAness.
Xenorhabdus and Photorhabdus bacteria are gaining more and more attention as a subject of research because of their unique yet similar life cycle with nematodes and insects. This work focused on the secondary metabolites that are produced by Xenorhabdus and Photorhabdus. With the help of modern HPLC-MS methodologies and increasingly available bacterial genome sequences, the structures of unknown secondary metabolites could be elucidated and thus their biosynthesis pathways could be proposed, too.
The first paper reported 17 depsipeptides termed xentrivalpeptides produced by the bacterium Xenorhabdus sp. 85816. Xentrivalpeptide A could be isolated from the bacterial culture as the main component. The structure of xentrivalpeptide A was elucidated by NMR and the Marfey´s method. The remaining xentrivalpeptides were exclusively identified by feeding experiments and MS fragmentation patterns.
The second paper described the discovery and isolation of xenoamicin A from Xenorhabdus mauleonii DSM17908. Additionally, other xenoamicin derivatives from Xenorhabdus doucetiae DSM17909 were analyzed by means of feeding experiments and MS fragmentation patterns. The xenoamicin biosynthesis gene cluster was identified in Xenorhabdus doucetiae DSM17909.
The manuscript for publication focused on the biosynthesis of anthraquinones in Photorhabdus luminescens. The Type II polyketide synthase for the biosynthesis of anthraquinone derivatives was discovered in P. luminescens in a previous publication by the Bode group,1 in which a partial reaction mechanism for the biosynthesis has been proposed. The manuscript reported in this thesis however elucidated the biosynthetic mechanisms in a greater detail as compared to the previous publication. Particularly, the biosynthetic mechanism was deciphered through heterologous expression of anthraquinone biosynthesis (ant) genes in E. coli. Additionally, deactivation of the genes antG encoding a putative CoA ligase and antI encoding a putative hydrolase, was performed in P. luminescens. Selected ant genes were over-expressed in E. coli as well as the corresponding proteins purified for in vitro assays. Model compounds were chemically synthesized as possible substrates of AntI and were used for in vitro assays. Here, it was revealed that the CoA ligase AntG played an essential role in the activation of the ACP AntF. Furthermore, a chain shortening mechanism by the hydrolase AntI was identified and was further confirmed by in vitro assays using model compounds. Additionally, this chain shortening mechanism was supported by homology based structural modeling of AntI.
The lung comprises more than 40 different cell types, from epithelial cells to resident mesenchymal cells. These cells arise from the foregut endoderm and differentiate into specialized cell types that form the respiratory and conducting airways, and the trachea. However, the molecular pathways underlying these differentiation processes are poorly understood, and may be relevant to pathological conditions. According to the World Health Organization (WHO), while the respiratory disease rate is increasing, limited treatment and therapies are available. Thus, there is a growing need for new treatment strategies and alternative therapies. Various in vivo and in vitro studies in the model organism mus musculus have already provided valuable information on lung cell lineages and their differentiation and/ or dedifferentiation during development and pathological conditions. However, there remain many questions regarding the key regulators and molecular machinery driving lung cell differentiation and underlying lung progenitor/stem cell biology.
Aiming to develop new animal models for lung diseases, we used a forward genetic careening approach, which provides an unbiased method for identifying genes with important roles in lung cell differentiation, and thus probable contributors to pathological conditions. We conducted an N-ethyl-N-nitrosourea (ENU) mutagenesis screen in mice and used several histological and immunohistochemical approaches to identify and isolate mutants, focusing on mutations associated with cell differentiation rather than those affecting early development and patterning of the respiratory system. Thus, we screened for phenotypes in the respiratory system of pups from the F2 generation at postnatal day 7 and 0 (P7; P0). I specifically screened 114 families. Each F1 male animal is the founder of 5 to 6 F2 female daughters. For each family, at least 4 F2 females per male founder were analyzed. In total, I screened 630 litters at P7 and P0 with 7 pups on average for each litter. As a result of this extensive screening, 11 different phenotypes in 42 different F2s were discovered at primary screen and later just 2 phenotypes recovered in F3 generation of identified carriers. To identify the causative genes for each of these phenotypes, whole exome sequencing will be conducted in the future to identify recurring SNPs; these can subsequently be linked causatively to the resultant phenotype(s) via complementation studies. In turn, these linkages would enable the creation of mutant mice using CRISPR/Cas9 genomic engineering, which would be invaluable to the further study of respiratory development and disease.
Juvenile neuronal ceroid-lipofuscinosis (JNCL) is a rare lysosomal storage disease in children with lethal outcome and no therapy. The origin of JNCL has been traced to autosomal recessive mutations in the CLN3 gene, and ~85% of the JNCL patients harbor a 1.02 kb deletion that removes the exons 7 and 8 and the surrounding intronic DNA (CLN3Δex7/8). So far, structure, function and localization of the CLN3 protein remain elusive. However, there is strong evidence that CLN3 modulates a process or condition that is essential in many cellular pathways. Lipid metabolism and antero-/retrograde transport, two mechanisms CLN3 was previously implicated in, fulfill these requirements. Notably, also a bioactive group of glycosphingolipids referred to as gangliosides is tightly interrelated with these functions. Furthermore, a-series gangliosides have been shown to be involved in the development and sustenance of the brain, where they are essential for neurite outgrowth and cell survival. Defects in ganglioside metabolism were shown to play a crucial role in many lysosomal storage disorders. However, the contribution of gangliosides to NCL pathology is largely unknown.
The present study analyzed central enzymes and metabolites of the a-series ganglioside pathway in a JNCL cell model. The core finding was, thereby, the reduced amount of the neuroprotective ganglioside GM1 in homozygous CbCln3Δex7/8 cells. This was caused by the enhanced action of the GM1-degrading multimeric enzyme complex and in particular, by the upregulation of protein levels and increased enzyme activity of β-galactosidase (Glb1).
Improved binding of Glb1 to substrate-carrying membranes was provided by an increase in LBPA levels. In combination with other smaller alterations in the ganglioside pattern, a shift towards less complex gangliosides became present. The resulting loss of neuroprotection may be the reason for the multifocal pathology in homozygous CbCln3Δex7/8 cells.
The second part of the present study investigated the cellular mechanisms behind the altered ganglioside profile with regard to the potential role of CLN3. Here, the anterograde transport of GM1 to the plasma membrane presented a positive correlation with the amount of full-length CLN3. In case of the truncated protein this correlation was missing, resulting in reduced PM staining with CTxB-FITC. However, transfection of full-length CLN3 in these cells restored the CTxB-FITC intensity. Based on the neuroprotective role of GM1, the corresponding increase in GM1 levels may be the cause for the restoration effects observed in previous studies using full-length CLN3. Hence, administration of GM1 was expected to improve cell viability of homozygous CbCln3Δex7/8 cells and beyond that to rescue potentially some disease phenotypes. However, no effect could be observed. The reason for this may be reduced caveolar uptake and the mislocalization of ganglioside GM1 to the trans-Golgi network (TGN) and redirection towards degradative compartments.
Both are in line with the idea of an impaired endocytic flux in CLN3 deficiency. The observed localization of CLN3 in the TGN suggests a potential role for CLN3 in the lipid sorting machinery, subsequently altering membrane composition and its regulatory functions. The resulting imbalance may affect many of the cellular processes impaired in JNCL.
Primäre Tumore werden nach ihrem Entstehungsort benannt. Selbst Metastasen zeigen eine gewisse Ähnlichkeit mit ihrem ursprünglichen Gewebe. Somit gehören alle Geschwülste, die ursprünglich der Harnblase entstammen, zu den Harnblasenkarzinomen.
Das Harnblasenkarzinom geht meist (90-95%) von der Schleimhaut der ableitenden Harnwege aus und wird als Urothel bezeichnet. Dementsprechend haben die meisten Patienten mit der Diagnose Blasenkarzinom ein Urothel-Blasenkarzinom. Die restlichen Blasenkarzinome entfallen auf Adenokarzinome, Plattenepithelkarzinome, kleinzellige Karzinome, Sarkome, Paragangliome, Melanome oder Lymphome (Humphrey A. et al. 2016, Moch H. et al 2016).
Die Urothel-Blasenkarzinome können sowohl flach, als auch warzenförmig wachsen. Je nach Diagnostik „oberflächlich“ oder „muskelinvasiv“ lassen sich die Urothel-Blasenkarzinome in zwei Hauptgruppen unterteilen;
Etwa 70% der Erkrankten haben dabei ein oberflächliches Urothel-Blasenkarzinom, das auf die Blasenschleimhaut begrenzt ist und durch eine Basistherapie, sog. Transurethrale Resektion (TUR-B) behandelt wird. Dabei werden die in der Schleimhaut gewachsenen Tumore getrennt reseziert. Therapieergänzend und/oder prophylaktisch wird die Blase danach mit einem Chemotherapeutikum gespült (intravesikale Instillation). Diese Zytostatika-Behandlung soll das Wiederauftreten eines Rezidivs verhindern bzw. eventuell verlangsamen (Iida K. et al. 2016, Celik O. et al. 2016).
Mitochondrial membrane dynamics is increasingly implicated in various human diseases. Numerous studies show that the protein OPA1 plays a central role in determining mitochondrial ultrastructure and apoptotic remodeling of the inner mitochondrial membrane during Cytochrome c release and apoptosis. Crista junctions are crucial for the regulation of apoptotic Cytochrome c release. Previous publications suggest that OPA1 is required to maintain a normal structure of the inner mitochondrial membrane. The protein MIC60 (Mitofilin) appears to be an essential physical constituent of crista junctions and is also crucial for the general determination of mitochondrial ultrastructure. Furthermore, recent studies suggest that MIC60 is also implicated in Cytochrome c release during apoptosis.
In this regard, the question whether OPA1 is essential for crista junction formation was investigated. In addition to that, the interplay between OPA1 and MIC60 and its physiological role were analyzed. Electron microscopy of OPA1+/- and OPA1+/+ mice, as well as of OPA1-/- and OPA1+/+ MEFs clearly showed that OPA1 plays a role but is not essential for crista junction formation. In contrast to that, the results indicate that OPA1 is crucial to maintain a normal structure of the inner mitochondrial membrane. Immunogold experiments fit well to these observations as OPA1 was found equally distributed throughout the cristae membrane with only a minor part located at crista junctions. MIC60 localization studies showed a clear enrichment at crista junctions. Interaction studies revealed that endogenous OPA1 and MIC60 physically interact with each other. Analysis of protein levels upon OPA1 or MIC60 depletion indicate that both proteins play a dual role in cristae- and crista junction formation in which MIC60 is a physical constituent of crista junctions essential for their formation while OPA1 primarily has a regulatory impact on MIC60 function. Finally, apoptosis assays and cell viability measurements showed that knockout of OPA1 in MEFs leads to increased cellular resistance suggesting that the interplay of these proteins is important for the regulation of crista junction remodeling during apoptosis.
Besides its role in determining mitochondrial ultrastructure, OPA1 mediates inner membrane fusion of mitochondria thereby contributing to mitochondrial quality control. Additionally, proteolytic processing is crucial for the ability of OPA1 to distinguish between functional and dysfunctional mitochondria. Functional mitochondria are fused while dysfunctional mitochondria are not, a process termed selective mitochondrial fusion. Dysfunctional mitochondria were shown to be degraded by mitophagy in a fission-dependent manner. Numerous studies suggest that OPA1 and mitophagy are directly linked. However, this idea is still under debate. Mitophagy is also crucial for mitochondrial quality control, which directly impacts mitochondrial integrity. Furthermore, mitochondrial quality control has been linked to neurodegeneration as demonstrated by the observation that mutations in OPA1 cause the disorder ADOA-1.
In order to analyze a potential link between OPA1 and mitophagy, mitochondrial colocalization with LC3 was analyzed microscopically in primary adult skin fibroblasts isolated from OPA1+/- and OPA1+/+ mice in an age-dependent manner. Fibroblasts from young OPA1+/- mice showed increased colocalization of mitochondria with autophagosomes compared to fibroblasts from young wild type mice suggesting that OPA1 exerts an inhibitory role in mitophagy. This effect was even more pronounced in old mice, which also displayed higher mitophagy levels in general than young mice, consistent with the finding that old mice had higher Parkin levels than young mice. Mitochondrial fragmentation was elevated in fibroblasts from young and old OPA1+/- mice compared to control fibroblasts. However, extensive mitochondrial fusion, which occurred in fibroblasts from old wild type mice, was prevented in old OPA1+/- mice. Furthermore, old wild type mice had decreased numbers of crista junctions compared to young wild type mice, an effect that was not observed in OPA1+/- mice. Despite the observed age-dependent phenotypes in mitochondrial quality control and mitochondrial integrity, deletion of one allele of OPA1 had no influence on the life span in vivo. Analysis of the OPA1-dependent proteome of aging mice, which was performed in collaboration with Ansgar Poetsch and Carina Ramallo-Guevara from Bochum, showed that OPA1-dependent aging is accompanied by a reduction of proteins involved in autophagy. In contrast to that, a switch from glucose to fatty acid metabolism and alterations in apoptotic proteins were observed in both OPA1+/- and OPA1+/+ mice in an age-dependent manner indicating that the changes in proteins implicated in autophagy could be a compensatory response to the diminished inhibitory effect of OPA1 on mitophagy. On the other hand, increased mitochondrial degradation by mitophagy could be a cellular response to itself compensating for the loss of OPA1 mediated fusion thereby contributing to the observation that OPA1+/- and OPA1+/+ mice had no differences in life span. Furthermore, analysis of the OPA1-dependent proteome of aging mice revealed that OPA1, besides its role in mitochondrial fusion, could interact with the fission machinery: MFF and Neuronal pentraxin 1, two proteins involved in mitochondrial fission, were up-regulated in 12-month-old OPA1+/- mice suggesting that a reduced fission activity could contribute to mitochondrial hyperfusion in aged wild- type mice. Nonetheless, the exact nature of the possible interplays between OPA1 and these candidates remains to be investigated.
Der Pilz Podospora anserina ist seit mehr als fünf Jahrzehnten ein wichtiger Modellorganismus für die Alternsforschung. Insbesondere die Mitochondrien, essentielle eukaryotische Zellorganellen – wegen ihrer Funktion im Energiestoffwechsel häufig auch als „zelluläre Kraftwerke“ bezeichnet, sind Schlüsselfaktoren für den Alterungsprozess dieses Organismus.
Im Rahmen einer vorangegangenen Diplomarbeit wurde daher der Einfluss der mitochondrialen CLPXP-Protease, einem bisher noch wenig erforschten Bestandteil der Proteinqualitätskontrolle in Mitochondrien, auf die Alterung von P. anserina untersucht. Mitochondriale CLPXP-Proteasen sind, wie auch ihre bakteriellen Pendants, aus zwei verschiedenen Untereinheiten aufgebaut: der Protease-Komponente CLPP und der Chaperon-Komponente CLPX. Die Deletion des Gens PaClpP, kodierend für CLPP in P. anserina, führte zu einer überraschenden Verlängerung der gesunden Lebensspanne der Mutante. Darüber hinaus war es möglich, den pilzlichen PaClpP-Deletionsstamm durch Einbringen von CLPP des Menschen zu komplementieren. Dies beweist, dass die Proteasen CLPP des Menschen und von P. anserina funktionell homolog sind. Dadurch eröffnete sich die Perspektive, diesen einfachen Modellorganismus für die Gewinnung potenziell auf den Menschen übertragbarer Erkenntnisse einzusetzen. Bedeutenderweise ist die menschliche CLPXP-Protease wahrscheinlich involviert in die Entstehung verschiedener Krankheiten, darunter das Perrault-Syndrom sowie einige Krebsarten. Die zugrundeliegenden Mechanismen sind jedoch noch weitestgehend unverstanden.
Ziel des in dieser Dissertation beschriebenen Forschungsprojektes war daher die Gewinnung genauerer Einsichten in die molekulare Funktion und die daraus folgende biologische Rolle der mitochondrialen CLPXP-Protease von P. anserina. Der wohl wichtigste Punkt für das detaillierte Verständnis einer Protease ist die Kenntnis ihres Substratspektrums, d. h. der von ihr abgebauten Proteine. Tatsächlich wurde aber bis heute noch in keinem eukaryotischen Organismus eine umfassende Analyse der Substrate einer mitochondrialen CLPXP-Protease vorgenommen. Um diese Wissenslücke zu füllen, wurde in der vorliegenden Arbeit eine ursprünglich in Bakterien entwickelte Verfahrensweise, der sogenannte CLPP „Substrat-trapping Assay“, in P. anserina implementiert. Dafür mussten zunächst die notwendigen handwerklichen Voraussetzungen für den Assay geschaffen werden, insbesondere die effiziente Affinitätsaufreinigung von Proteinen aus isolierten Mitochondrien – einer bisher in P. anserina noch nicht angewandten Technik. Unter Verwendung verschiedener neu hergestellter Varianten der menschlichen Protease-Komponente CLPP, darunter einer proteolytisch inaktiven Variante zum „Einfangen“ von Substraten, konnte der CLPP „Substrat-trapping Assay“ in P. anserina erfolgreich durchgeführt werden. Insgesamt wurden, in Kooperation mit der Arbeitsgruppe von Julian D. Langer (Max-Planck-Institut für Biophysik; Durchführung von massenspektrometrischen Analysen) nahezu 70 spezifische Proteine erstmalig als potenzielle Substrate oder Interaktionspartner einer mitochondrialen CLPXP-Protease identifiziert. Bei einem Großteil dieser Proteine handelt es sich um Enzyme und Komponenten verschiedener Stoffwechselwege – vor allem um solche, die eine zentrale Rolle im mitochondrialen Energiestoffwechsel spielen. Die Ergebnisse der vorliegenden Arbeit legen somit folgende Arbeitsthese als Schlussfazit und gleichzeitig Ausganspunkt für zukünftige Untersuchungen nahe:
Die hauptsächliche molekulare Funktion der mitochondrialen CLPXP-Protease in P. anserina ist die Degradation von Stoffwechselenzymen und ihre biologische Rolle demnach die Kontrolle und Aufrechterhaltung des mitochondrialen und zellulären Energiestoffwechsels.
Insgesamt ist die auf Grundlage des CLPP „Substrat-trapping Assay“ in P. anserina anzunehmende Rolle der mitochondrialen CLPXP-Protease als regulatorische Komponente des mitochondrialen Energiestoffwechsels erstaunlich gut mit Beobachtungen in anderen eukaryotischen Organismen, gerade bezüglich der Relevanz der CLPXP-Protease des Menschen für diverse Krankheiten, zu vereinbaren. Somit erscheint es überaus sinnvoll und vielversprechend, dass in dieser Doktorarbeit erstellte und bisher beispiellose Kompendium potenzieller in vivo Substrate und Interaktionspartner dieser Protease auch als Referenz für zukünftige Untersuchungen außerhalb von P. anserina anzuwenden.
Reading is an essential ability to master everyday life in our society. The ability to read is based on specific connections between brain regions involved in the reading process – so-called cortical networks for reading. These cortical networks for reading allow us to learn the correct identification of visual words. The use of visual words is based on knowledge about the orthography (lexical) and the meaning of words (semantic). This knowledge must be acquired by beginning readers (first grader), i.e. beginning readers learn in a first step to link letters to a whole word and in a second step associate this whole word with meaning. To retrieve this knowledge during visual word recognition (VWR) a cortical network for lexical-semantic process must be activated. However, it is currently unclear whether beginning readers and reading experts activate the same neuronal network during VWR. Therefore, the aim of this thesis was to investigate the question whether beginning readers (first grader, children) and reading experts (adults) use different cortical networks for the lexical-semantic processing in VWR.
To address this question we recorded electroencephalographic (EEG) activity during VWR in children and adults. Children and adults were instructed to read a visualizable word to compare this word with a following picture stimulus. The first part of this thesis is concerned with the analysis of ERPs for visual word recognition in children and adults at sensor level. For both groups we observed the typical ERP components P100 and N170 for visual word recognition. These components differed in amplitude and time course between both groups. The second part of this thesis investigated the neuronal generators (brain areas) of ERPs during VWR and possible differences between children and adults at source level. We observed a high overlap in brain areas involved during VWR in children and adults. However, the brain areas differed in activation and time course between children and adults. Finally, the third and most important part of the thesis investigated the question whether children and adults use different cortical networks for the lexical-semantic processing in VWR over time. To address this question Dynamic Causal Modeling (DCM) and Bayesian model comparison were used. We compared nine biologically plausible cortical network models underlying the ventral lexical-semantic path in VWR. In addition, increasing time intervals were used to consider possible changes of network structure during VWR. The network models included eight brain regions (four bilateral pairs) involved in the lexical-semantic processing in VWR: occipital cortex (OC), temporo-occipital part of inferior temporal gyrus (ITG), temporal pole (TP), and inferior frontal gyrus (IFG). In almost all time intervals we found evidence that children and adults use the same cortical networks for the lexical-semantic processing in VWR. However, we found differences between adults and children in the connection strengths of the favoured model. Interestingly, we found a stronger direct connection from OC to IFG in adults compared to children.
In conclusion, our results suggest that children and adults activate largely the same lexical-semantic networks during VWR over time. This supports the notion that children and adults use the same biological fiber connections for VWR. However in contrast to children, adults showed increased use of the shortcut pathway from OC to IFG. The increased use of the shortcut pathway from OC to IFG in adults can be interpreted as consequence of learning. Learning causes in accordance with the Hebbian learning rule (“neurons that fire together, wire together” (Hebb, 1949)) synaptic change. Consequently the frequent coactivation of the input and output stage of OC and IFG during the lexical-semantic process facilitates the stronger direct connection between both brain areas. The stronger direct connection from OC to IFG most likely allows adult reading experts to speed up the lexical-semantic process during VWR. Accordingly, we conclude that the stronger direct connections from OC to IFG in adults compared to children underlay the different reading capabilities in both groups.
Identification of disease modulating compounds in juvenile neuronal ceroid lipofuscinosis (JNCL)

Mutationen im CLN3 Gen verursachen die neurodegenerative Erkrankung juvenile neuronale Zeroidlipofuszinose (JNCL). Bei dieser Erkrankung sind die Autophagie, der lysosomale pH Wert und der mitochondriale Metabolismus beeinträchtigt. Störungen dieser Prozesse führen zu einer erhöhten Verletzlichkeit neuronaler Zellen gegenüber alters- und umweltbedingten Schäden, einer Anhäufung von Autophagosomen und lysosomalem Speichermaterial, Zelltod und Neurodegeneration. Um die JNCL zu erforschen bedienen wir uns eines Zellmodels aus der Maus, welches die häufigste krankheitsauslösende CLN3 Mutation im Menschen, die Deletion der Exons 7 und 8, nachbildet. Die aus dem Kleinhirn dieser Mäuse stammenden cerebellaren Körnerstammzellen werden als CbCln3Δex7/8/Δex7/8 Zellen, solche aus wild-typ Mäusen als CbCln3+/+ Zellen bezeichnet. Die JNCL ist nicht heilbar und die Entwicklung von Wirkstoffen steht noch am Anfang.
Die vorliegende Arbeit befasst sich mit der Durchführung eines Hochdursatzscreenings um Wirkstoffe zu identifizieren, welche eine Anhäufung von Autophagosomen in CbCln3Δex7/8/Δex7/8 Zellen verhindern können. Unter 1750 verschiedenen untersuchten Wirkstoffen konnten wir 28 aktive „Hits“ identifizieren und stellten fest, dass Kalziumkanalblocker, Östrogene und HMG-CoA-Reduktase Inhibitoren gehäuft vertreten waren. Eine sorgfältige Untersuchung die möglichen Interaktionen der aktiven Wirkstoffe mit zellulären Signalwegen und die Analyse ihrer Dosis-Wirkungskurven unterstützte uns bei der Auswahl von Verapamil, Nicardipin und Fluspirilen zur näheren Untersuchung. Diese Wirkstoffe sind Kalziumkanalblocker und Fluspirilen blockt auch D2 Dopaminrezeptoren.
Außerdem untersuchten und quantifizierten wir mitochondriale Phänotypen in CbCln3Δex7/8/Δex7/8 Zellen. Unsere Untersuchungen ergaben, dass Mitochondrien in CbCln3Δex7/8/Δex7/8 Zellen einer signifikanten Hyperfusion unterliegen und ein schwächeres Membranpotenzial aufweisen. Weiterhin fanden wir eine Verringerung der maximalen der mitochondrialen Elektronentransportkapazität und eine verringerte Aktivität des Enzyms Zitratsynthase, welches die Effizienz des Zitratzyklus bestimmt.
Fluspirilen, Verapamil und, in geringerem Ausmaß, Nicardipin, verbesserten einige krankheitsbedingte lysosomale und mitochondriale Phänotypen. Des Weiteren konnten Verapamil und Nicardipin, nicht aber Fluspirilen, den erhöhten zellulären Kalziumspiegel in CbCln3Δex7/8/Δex7/8 Zellen absenken. Erniedrigungen im Kalziumgehalt können durch die Inhibition der kalziumabhängigen Protease Calpain 1 zu einer Induktion der Autophagie führen. Wir untersuchten, ob eine chemische Inhibition der Calpain 1-Protease die Anzahl der Autophagosomen in CbCln3Δex7/8/Δex7/8 Zellen senkt, und stellten fest, dass dies nicht der Fall ist. Eine Inhibition von Calpain 1 führte lediglich zu einem Anstieg der Zahl zellulärer Autophagosomen. Als Nächstes untersuchten wir die Auswirkung der Wirkstoffbehandlung auf den Autophagiefluss. Verapamil und Nicardipin hatten keinen Einfluss auf den Autophagiefluss in der getesteten Konzentration in CbCln3Δex7/8/Δex7/8 Zellen während Fluspirilen die Autophagie induzierte. Gleichzeitig stellten wir fest, dass hohe Dosen von Nicardipin und Verapamil teilweise vor einem Verlust des lysosomalen pH-Werts durch eine Behandlung mit Bafilomycin A1 schützen konnten. Da Fluspirilen auch ein Dopaminrezeptorblocker ist, untersuchten wir die Auswirkung einer erhöhten Dosis von Dopamin auf die Zahl der Autophagosomen. Wir fanden, dass eine mittlere Dosierung von Dopamin einen Trend zu einer leichten Verringerung von Autophagosomen in CbCln3Δex7/8/Δex7/8 Zellen zur Folge hat.
Wir vermuten, dass die Kalziumkanalblocker Verapamil und Nicardipin und der Dopaminrezeptorblocker Fluspirilen unterschiedliche zelluläre Signalwege benutzen, aber letztendlich um ähnliche Botenstoffe verwenden, um die Funktion der Lysosomen in CbCln3Δex7/8/Δex7/8 Zellen zu verbessern. Die Verringerung des intrazellulären Kalziumgehalts durch Verapamil und Nicardipin führt zu einer Aktivierung von Adenylatzyklasen, welche eine Erhöhung des intrazellulären cAMP Spiegels herbeiführen. Fluspirilen inhibiert Dopaminrezeptoren vom Typ D2 (D2DR), was zu einer selektiven Aktivierung von Dopaminrezeptoren des Typs D5 (D5DR) führen könnte. Im Gegensatz zu D2 führen D5D Rezeptoren zu einer Aktivierung von Adenylatzyklasen und einer Erhöhung des cAMP Spiegels. cAMP aktiviert die Protein Kinase A (PKA), welche durch eine Proteinphosphorylierung von lysosomalen Chloridkanälen und Protonenpumpen die lysosomale Aktivität erhöht. Dies führt zu einer Verbesserung des Abbaus von Autophagosomen und lysosomalem Speichermaterial und zu einer verbesserten Zellgesundheit in CbCln3Δex7/8/Δex7/8 Zellen.
Eine Verbesserung der lysosomalen Funktion in der JNCL kann einen wirksamen Therapieansatz ergeben. Wir hoffen, dass die hier vorgestellten Methoden und Ergebnisse einen ersten Schritt in diese Richtung darstellen.
Das Hören hat für den Menschen eine maßgebliche Bedeutung hinsichtlich Kommunikation und Orientierung. Auch wenn sich der Mensch stark auf seinen visuellen Sinn verlässt, wird mit dem Ausfall des Hörvermögens deutlich, wie viele Informationen oft unterbewusst über die Analyse von Schallsignalen gezogen werden. Trotz dieser grundlegenden Relevanz sind bis heute noch nicht alle Komponenten, die dem Hörprozess zugrunde liegen, entschlüsselt.
Um sich diesen offenen Fragestellungen anzunähern, müssen Forscher oft auf Tiermodelle zurückgreifen. Auf Grund ihres exzellenten Gehörs haben sich hier in den letzten Jahrzehnten Fledermäuse als taugliche Versuchstiere qualifiziert. Diese Tiere sind in der Lage sich ohne Verwendung des visuellen Systems in absoluter Dunkelheit zu orientieren, indem sie mit Hilfe der wiederkehrenden Echos ihrer ausgesendeten Ultraschalllaute die Umgebungsstrukturen analysieren. Weiterhin umfasst der zur Kommunikation und Ortung verwendete Frequenzbereich bei Fledermäusen ein Vielfaches von dem des menschlichen, was ebenfalls verschiedene Aspekte der Hörforschung begünstigt. Die in dieser Studie verwendete fruchtfressende Fledermausart Carollia perspicillata eignet sich hervorragend für akustische Untersuchungen, da ihr Innenohr keine speziellen morphologischen Spezialisierungen aufweist.
Anhand der Fledermausart C. perspicillata sollen innerhalb der vorliegenden Studie verschiedene offene Fragestellungen bezüglich der Innenohrmechanik näher beleuchtet werden. Um sich diesen Fragestellungen anzunähern, wurde eine Kombination aus zwei etablierten Methoden verwendet. Zum einen die Messung von Distortions-Produkt otoakustischen Emissionen (DPOAEs), welche auf Grund ihrer Generierung durch aktive Prozesse innerhalb der Kochlea die Möglichkeit bietet, Veränderungen im Innenohr festzustellen und zum anderen kontralaterale akustische Stimulation (KAS), welche eine erprobte Methode zur Aktivierung des efferenten Systems darstellt. Dadurch, dass die äußeren Haarsinneszellen in der Kochlea direkte synaptische Kontakte mit efferenten Fasern der absteigenden Hörbahn eingehen, kann eine Aktivierung des efferenten Systems Modulationen des kochleären Verstärkers bewirken, wodurch sich wiederum die Antworteigenschaften der Kochlea verändern. Mit einer Kombination dieser beiden Methoden lassen sich demnach zum einen höhere Zentren der Hörbahn aktivieren, die über efferente Fasern einen direkten Einfluss auf das Innenohr nehmen, zum anderen die induzierten Modulationen in Form von DPOAEs mit Hilfe eines sensitiven Mikrofons aufnehmen. Die Grundvoraussetzung für die Funktionalität dieser Methodenkombination ist das Vorhandensein von efferenten Fasern innerhalb der Kochlea. Da das efferente System verschiedener Säuger eine große Diversität aufweist, wurden innerhalb dieser Arbeit zusätzlich zu den akustischen Untersuchungen histologische Schnittserien der Kochlea von C. perspicillata angefertigt. Hierbei lag das Hauptaugenmerk auf dem Verlauf der efferenten Fasern innerhalb der Kochlea. Mit Hilfe der Thiocholinmethode wurde der Ort der Umsetzung des Achetylcholinabbauenden Enzyms Achetylcholin-esterase angefärbt. Achetylcholin ist der vorranig vorkommende Transmitter an den efferenten Synapsen.
Diese Studie untersucht weiter den Einfluss der Narkose auf das Innenohr. In zahlreichen Studien, die Innenohrmechanik betreffend, wurden die Untersuchungen an narkotisierten Tieren durchgeführt. Oftmals wird zwar die Problematik der möglichen Beeinflussung des Innenohres durch das verwendete Narkosemittel diskutiert, aber meisthin als unumgänglich eingestuft. Innerhalb der vorliegenden Studie wurde ein Großteil der Experimente an narkotisierten und auch wachen Tieren durchgeführt, um die Auswirkungen der häufig verwendeten Ketamin-Xylazin-Narkose auf die Innenohr-aktivität zu verdeutlichen.
In der Literatur lässt sich eine Vielzahl von akustischen Untersuchungen finden, in denen artifizielle Stimuli wie Reintöne oder Rauschen verwendet werden. Die Problematik dahinter ergibt sich daraus, dass diese Art der Töne in der Natur selten zu finden sind. Derartige Studien werfen daher die Frage auf, ob das Innenohr beispielsweise Rauschstimuli auf die gleiche Weise verarbeitet wie natürliche, komplexere Stimuli. Innerhalb der vorliegenden Studie wurden demzufolge im Vergleich zu artifiziellen Stimuli arteigene Rufe der Fledermausspezies C. perspicillata aufgenommen und als akustische Stimuli während der Messungen verwendet.
Im Zuge dieser Fragestellung wurde in einem weiteren Teilprojekt versucht ein neues Verfahren zur Messung von OAEs zu etablieren, mit dem es möglich ist das Ohr nicht ausschließlich mit den herkömmlich verwendeten Reintönen zu stimulieren, sondern ebenfalls mit komplexen Lauten, wie arteigenen Kommunikations- und Echo-ortungsrufen. Hierfür wurde ein von Douglas Keefe vorgestelltes Paradigma zur Messung von OAE-Residualen herangezogen, welches die am Trommelfell gemessenen akustischen Signale von den Trommelfellantworten auf einzelne Komponenten dieser Signale subtrahiert.
Anhand der in dieser Studie gewonnenen Ergebnisse kann deutlich gezeigt werden, dass eine akustische Stimulation der kontralateralen Kochlea mit verschiedenen artifiziellen sowie arteigenen Stimuli zuverlässig eine Änderung des Pegels der 2f1-f2 DPOAE von bis zu 37,3 dB in der ipsilateralen Kochlea bei wachen Tieren bewirkt. Dabei unterscheidet sich die Art der Beeinflussung deutlich je nach verwendetem kontralateralem Stimulus. Die Stimulation mit artifiziellem Breitbandrauschen supprimiert den Emissionspegel über den gesamten getesteten Frequenzbereich um etwa 11,6 dB, während die verwendeten arteigenen Laute eine vergleichbare Beeinflussung des DPOAE-Pegels ausschließlich in einem Frequenzbereich zwischen 50 und 70 kHz bewirken. Im Frequenzbereich von 20 bis 30 kHz verursachen die arteigenen Laute nahezu keine Pegelabsenkung, was im deutlichen Kontrast zu den Ergebnissen unter KAS mit Breitbandrauschen steht. Unter Narkoseeinfluss konnte, unabhängig vom verwendeten Stimulus, keine Beeinflussung des DPOAE-Pegels festgestellt werden, was die Annahme bestätigt, dass die verwendete Ketamin-Xylazin-Narkose einen drastischen Einfluss auf den Hörprozess und insbesondere auf das efferente System nimmt. Die Ursache dafür, dass arteigene Stimuli anders verarbeitet werden als artifizielle Stimuli (wie z. B. Breitbandrauschen) konnte zwar nicht abschließend geklärt werden, aber die Vermutung liegt nahe, dass in diesem Verarbeitungsprozess höhere Zentren der Hörbahn involviert sind und selektiven Einfluss auf die ablaufenden Prozesse nehmen.
Die Etablierung des OAE-Residual-Messparadigmas auf der Basis der Methode von Keefe und Ling (1998) sollte die Möglichkeit bieten sowohl Reintöne als auch komplexe, arteigene Stimuli zu verwenden und so eine Erweiterung des herkömmlich verwendeten Messverfahrens darstellen. Über verschiedene Vorversuche unter Anwendung einer Zweitonreizung konnte gezeigt werden, dass die Ergebnisse des neu entwickelten Paradigmas mit denen der herkömmlichen DPOAE-Messungen hinsichtlich Reintonstimuli vergleichbar sind. Anhand der gewonnenen Ergebnisse mit einer Stimulation mit komplexen Signalen zeigen sich allerdings die Schwierigkeiten der neuen Methode. Die bisher erhobenen Daten zeigen keine klaren, reproduzierbare Ergebnisse, sollten aber die Grundbedingungen für die weiterführenden Versuche ebnen.
Rhythms, i.e. periodic sequences of events or states, are a ubiquitous feature of physiological systems such as the heart, the lungs or the brain. For the brain in particular, the diversity of rhythms is remarkable, ranging from low frequency rhythms in the slow/delta band (0.5-4 Hz) during sleep to gamma band oscillations (30-120 Hz) rhythms during alert behavior, all expressed in various brain areas and at various spatial scales. To understand whether these rhythms subserve a function for the organism it is important to also understand the underlying mechanisms that generate them. While the generation of some rhythms appear to be well-understood, e.g. sleep spindles, others such as the cortical beta rhythm (13-30 Hz) have remained elusive.
Understanding the generation of a brain rhythm involves multiple spatial scales, from identifying intracellular mechanisms such as the contribution of individual transmembrane currents to studying how specific neuronal populations or areas affect the full physiological rhythm present in the intact, highly interconnected brain. The aim of this work has been to delineate the mechanistic contributions of individual brain areas to the in vivo generation of two particular rhythms present in efferent areas: (1) The first part of this work studies the influence of thalamocortical neurons on cortical slow/delta waves (0.5-4 Hz) of sleep that are sometimes also present in awake animals. (2) The second part is about the contribution of primary visual cortex to the beta rhythm (13-30 Hz) in extrastriate cortex of awake behaving animals.
Mikroalgen wird aufgrund ihrer photoautotrophen Lebensweise, ihrer meist einfachen Anzucht und ihres schnellen Wachstums ein großes Potential als Produzenten verschiedener Stoffe, wie beispielsweise den Sekundärmetaboliten der Carotinoidbiosynthese, zugesprochen. Zur Produktion solcher Stoffe bedarf es der Aufklärung der in einem Biosyntheseweg operierenden Enzyme und ihrer zugehörigen Gene.
In dieser Arbeit sollte einerseits durch genetische Modifikation der Carotinoidbiosynthese der Fucoxanthingehalt erhöht und andererseits die Produktion von Astaxanthin in P. tricornutum erreicht werden. Bisher fehlen experimentelle Nachweise über die Funktion, Regulation und die limitierenden Eigenschaften daran beteiligter Gene und deren Enzyme. Um dem Ziel der Arbeit näher zu kommen, wurden zuerst potentiell an der Regulation der Carotinoidbiosynthese beteiligte Gene ausgewählt und deren Enzyme funktionell charakterisiert. Eines dieser Enzyme ist das Eingangsenzym der Carotinoidbiosynthese, die Phytoen-Synthase. Die entsprechend annotierte putative Sequenz (psy #Pt56881) wurde zur Analyse herangezogen. Nachdem im Rahmen dieser Arbeit über die Komplementation in einem dafür ausgerichteten E. coli Stamm der funktionelle Nachweis der Phytoen-Synthase erbracht werden konnte, wurde untersucht, ob die Phytoen-Synthase einer lichtabhängigen Expression unterliegt und somit die Carotinoidsynthese im WT von P. tricornutum limitiert. Durch die Inhibierung der Phytoen-Desaturase mittels Norflurazon konnte die verstärkte Akkumulation des Produktes der Phytoen-Synthase, Phytoen, bei einem Transfer der P. tricornutum-Kulturen von Schwach- in Starklicht gezeigt werden. Die Expression der Phytoen-Synthase von P. tricornutum wird demnach durch die Lichtbedingungen reguliert und limitiert auch die Carotinoidsynthese. Ein weiteres an der Carotinoidsynthese beteiligtes Enzym ist die Zeaxanthin-Epoxidase. Sie bietet zugleich eine Möglichkeit, an dieser Stelle die Carotinoidsynthese in Richtung Astaxanthinproduktion umzulenken. Für P. tricornutum sind drei potentielle Genkandidaten (zep1: #Pt45845; zep2: #Pt56488; zep3: #Pt56792) annotiert, welche ebenfalls im Rahmen dieser Arbeit funktionell charakterisiert wurden. Der funktionelle Nachweis erfolgte dabei ebenfalls mittels eines Komplementationsansatzes in einem damit neu etablierten Expressionssystem mit npq2-Mutanten aus der Modellpflanze A. thaliana. Die Analyse der Transformanden zeigte eine Epoxidase-Aktivität des Produktes aus zep2 und zep3. Das Enzym Zeaxanthin-Epoxidase 2 weist dabei eine andere Spezifität auf als die Zeaxanthin-Epoxidase 3, welche funktionell betrachtet der Zeaxanthin-Epoxidase aus A. thaliana am nächsten kommt. Die Zeaxanthin-Epoxidase 2 akzeptiert im Unterschied zu Zeaxanthin-Epoxidase 3 neben Zeaxanthin auch andere Substrate wie Lutein mit nur einem 3 Hydroxy-β-Iononring und stellt damit einen validen Kandidaten für die Umwandlung von Diatoxanthin in Diadinoxanthin in P. tricornutum dar. Obwohl die Transkriptanalysen ausreichende Mengen an RNA von zep1 in A. thaliana zeigen und anhand eines zusätzlichen mit der Sequenz für GFP markierten zep1-Konstruktes in WT-Protoplasten von A. thaliana der Import in den Chloroplasten und die Expression nachgewiesen werden konnte, weist die Zeaxanthin-Epoxidase 1 zumindest in den A. thaliana-Transformanden keine Epoxidase-Aktivität auf. Des Weiteren zeigt die diurnale Expression in P. tricornutum, dass die Regulation der Zeaxanthin-Epoxidasen an den Bedarf photoprotektiver Pigmente angepasst wird. Während die Regulation des Transkript-Levels von zep2 und zep3 nahezu parallel laufen und ein gemeinsames Maximum aufweisen, zeigt das Transkript-Level von zep1 ein anderes Maximum.
Die gewonnenen Erkenntnisse wurden dann zur Steigerung der Synthesekapazität mittels genetischer Modifikation des Carotinoidsyntheseweges in P. tricornutum angewendet. Durch das Einbringen zusätzlicher Genkopien der Phytoen-Synthase in P. tricornutum konnte dabei eine deutliche Steigerung des Fucoxanthingehalts unter Schwachlichtbedingungen erreicht werden. Gleichzeitig konnte durch weitere inhibitorische Versuche mittels Norflurazon beim Transfer von Schwach- zu Starklicht demonstriert werden, dass die Carotinoidsynthese durch die Kombination der genetischen Modifikation mit der Phytoen-Synthase und Starklicht per se weiterhin gesteigert werden kann. Zusammen mit den Transkriptanalysen zeigen die Pigmentanalysen, dass es einen nicht-linearen Zusammenhang zwischen RNA-Menge und gebildeter Phytoenmenge gibt, welcher durch eine zusätzliche Substratlimitierung der Phytoen-Synthase erklärt werden kann.
Bevor das Herunterregulieren der Zeaxanthin-Epoxidasen in P. tricornutum durchgeführt und damit ein verstärkter Fluss zur Astaxanthinbildung erreicht werden sollte, wurde das Potential von P. tricornutum zur Astaxanthinproduktion überprüft. Hierfür wurde die β-Carotin-Ketolase (bkt #CrAEA35045.1) aus C. reinhardtii einmal ohne und zusätzlich mit verschiedenen Präsequenzen fusioniert separat in P. tricornutum eingebracht. Astaxanthin konnte trotz Nutzung funktionell bestätigter Präsequenzen aus der Literatur nicht nachgewiesen werden. Die Versuche zeigen damit, dass hier noch weitere Untersuchungen nötig sind, um mittels eines geeigneten Transportsystems Fremd-Proteine in den Chloroplasten von P. tricornutum einzubringen. Das Ausbleiben der Astaxanthinproduktion konnte an dieser Stelle nicht hinreichend geklärt werden.
Insgesamt schaffen die Ergebnisse dieser Arbeit eine weitere Grundlage, um die Carotinoidbiosynthese in P. tricornutum besser zu verstehen und diese mittels genetischer Modifikationen biotechnologisch nutzbar zu machen.
Diese Dissertation befasst sich mit den Auswirkungen von nicht letalen Dosen von Neonikotinoiden auf Bienen. Neonikotinoide stellen eine Klasse von Insektiziden dar, die auf den nikotinischen Acetylcholin Rezeptor wirken. In dieser Dissertation wurden die Neonikotinoide Imidacloprid, Clothianidin und Thiacloprid benutzt. Die beiden erst genannten unterliegen zum Zeitpunkt des Verfassens dieser Arbeit einem temporären Verkaufs- und Ausbringungs-Stopp. Damit sind die Ergebnisse dieser Arbeit wichtig für die Bewertung der Gefahren von Neonikotinoiden. Neonikotinoide werden im großen Maße in der Landwirtschaft als Spritzmittel und Saatgutbeize eingesetzt. Dabei können sie in Rückständen von Bienen beim Sammeln von Nektar und Pollen aufgenommen und zum Stock gebracht werden. Um einen weiten Blick auf die Auswirkungen der Stoffe zu werfen wurden deshalb Experimente an einzelnen Sammlerinnen durchgeführt, ebenso wie an Bienenvölkern, bei denen die Substanzen verfüttert wurden. Als neuronal aktive Substanzen können sie die normale Funktion des Nervensystems von Bienen beeinflussen, was Veränderungen im Verhalten hervorrufen kann. Dies zeigt sich in Veränderungen in der Bewegung, Orientierung oder auch Interaktion mit anderen Bienen. Die Wirkung am Rezeptor variiert, trotz gleichen molekularen Ziels, stark zwischen den verwendeten Neonikotinoiden. Clothianidin wurde als Agonist beschrieben, der sogar stärkere Ströme als Acetylcholin bei gleicher Konzentration hervorrufen kann. Imidacloprid dagegen wurde bereits als partieller Agonist beschrieben, der geringere Ströme über den Rezeptor auslöst. In dieser Arbeit wurde ein erster Versuch durchgeführt um Thiacloprid ebenfalls als Agonist am nikotinischen Acetylcholin Rezeptor der Biene zu beschreiben. Hierbei wurde an einer Zelle in Kultur ein geringerer Strom ausgelöst.
Bienenvölker wurden unter kontrollierten Bedingungen gehalten, bei denen je eins der Neonikotinoide Clothianidin, Imidacloprid oder Thiacloprid in das Futter gemischt wurden. Hierfür wurden Dosen gewählt, bei denen davon ausgegangen werden konnte, dass keine akute Beeinflussung der Sammlerinnen bestand. Es konnte festgestellt werden, dass chronisches Füttern mit einer Zuckerlösung mit 8,876 mg/kg Thiacloprid zu einer verringerten Sammelleistung führte. Ebenso wurde die Entwicklung der Eier stark eingeschränkt, wobei die Königin weiterhin Eier legte. Es konnten nur vereinzelte verdeckelte Brutzellen, die ein spätes Entwicklungsstadium der Bienen darstellen, gefunden werden. Damit konnte gezeigt werden, dass geringe Dosen die Larval-Entwicklung von Bienen beeinflussen, eventuell durch Einflüsse auf die Kommunikation zwischen Ammenbienen und der Brut.
Um Auswirkungen auf einzelne Tiere zu zeigen, wurden unterschiedliche Parameter im Heimflug von Bienen nach Fütterung mit je einem der Neonikotinoide analysiert. Bienen mussten sich nach der Fütterung orientieren und von einer neuen Position den Heimweg zum Stock finden. Der Heimflug wurde per Radar verfolgt und so ein Flugprofil erstellt, das aus zwei Flugphasen bestand. Diese wurden durch die Navigation nach Vektorintegration und durch Landmarken unterteilt. Aus dem Flugprofil konnte abgelesen werden, wie lange die Bienen für die Phasen des Flugs benötigten, in welchem Hauptflugwinkel sie die erste Flugphase absolvierten, in welche Richtung sie am Ende der ersten Flugphase flogen und wie gerichtet der Flug war. Auch wurde erfasst, ob die Bienen überhaupt in der Lage waren zum Stock zurückzukehren. Hier zeigte sich, dass die Fütterung mit Zuckerwasser mit 0,6 µM und 0,9 µM Imidacloprid, ebenso wie mit 0,1 mM Thiacloprid zu einer verringerten Heimkehrwahrscheinlichkeit führte. In der ersten Flugphase konnte auch gezeigt werden, dass 0,2 µM Clothianidin im Zuckerwasser zu einem schnelleren Flug führte und dass der Flugwinkel im Vergleich zur Kontrolle in Richtung der wahren Position des Stocks verschoben war. Beide Imidacloprid-Gruppen zeigten eine ähnliche, signifikante Verschiebung des Flugwinkels, ebenso konnte im Flug selbst eine häufige Änderung der Richtung festgestellt werden. In der zweiten Flugphase zeigte sich, dass Bienen, welche mit Thiacloprid behandelt wurden häufiger eine inkorrekte Heimflugrichtung wählten, was in längeren Heimflügen resultierte. Die mit Clothianidin behandelten Bienen legten eine längere Flugstrecke zurück. Bienen, welche Imidacloprid beider Konzentrationen konsumierten, zeigten einen häufigen Wechsel ihrer Flugrichtung. Damit konnten bei allen drei gewählten Neonikotinoiden Einflüsse auf spezifische Komponenten der Navigation von Bienen gefunden und Einschränkungen im Heimkehr- und Orientierungsverhalten einzelner Sammlerinnen gezeigt werden. Somit konnten die eingehenden Fragen zumindest teilweise beantwortet werden und die Datenlage zur Frage der Schädlichkeit der, auch politisch umstrittenen, Substanzen erweitert werden.
Flow hemodynamics regulates endothelial cell (EC) responses and laminar shear stress induces an atheroprotective and quiescent phenotype. The flow-responsive transcription factor KLF2 is a pivotal mediator of endothelial quiescence, but the precise mechanism is unclear. In this doctoral study, we assessed the hypothesis that laminar shear stress and KLF2 regulate endothelial quiescence by controlling endothelial metabolism.
Laminar flow exposure and KLF2 over expression in HUVECs reduced glucose uptake. Endothelial specific deletion of KLF2 (EC-KO) in mice and subsequent infusion of labeled glucose in Langendorff perfused hearts induced glucose uptake in ECs lacking KLF2. Bioenergetic measurements revealed that KLF2 reduces and glycolytic acidification in vitro.
Mechanistically, RNA sequencing analysis of shear stimulated ECs showed reduced expression of key glycolytic enzymes Hexokinase 2, PFKFB3 and PFK-1. KLF2 also reduced expression of these enzymes at protein level. KLF2 knockdown in shear stimulated ECs reversed the reduction in expression of PFKFB3 and PFK-1, indicating KLF2-dependency. Promoter analysis revealed KLF binding sites in the promoter of PFKFB3 and KLF2 over expression markedly reduced PFKFB3 promoter activity which was abolished on mutation of the KLF binding site. In addition, PFKFB3 knockdown reduced glycolysis while over expression increased glycolysis. Over expression of PFKFB3 along with KLF2 partially reversed the KLF2-mediated reduction in glycolysis. Importantly, PFKFB3 over expression reversed KLF2-mediated reduction in angiogenic sprouting and network formation in vitro. Ex-vivo aortic ring assays revealed an increase in endothelial sprouting from aortas from KLF2 EC-KO mice, which was partially reversed upon PFKFB3 inhibition by 3-PO.
In conclusion, work performed during this doctoral thesis demonstrates that laminar shear stress and KLF2 mediated repression of endothelial metabolism via regulation of PFKFB3 contributes to the anti-angiogenic and quiescent properties of the endothelium.
Dissecting the complexities of mammalian heart development and regenerative capacity require thorough understanding of the underlying molecular mechanisms through the expression pattern of proteins and post-translational modifications. To obtain insights intoactivated signaling pathways that control the cellular phenotype during postnatal heart development, we generated a comprehensive map of phosphorylation sites. In total we identified 21,261 phosphorylation sites and 8985 proteins in developing mouse hearts by mass spectrometry. The in-vivo SILAC (stable isotope labeling of amino acids in cell culture) approach allowed robust quantification of phosphorylation sites and proteins, which are regulated during heart development. We found several activated pathways involved in cell cycle regulation and detected numerous kinases and transcription factors to be regulated on protein and phosphopeptide level. Most strikingly, we identified a novel mitochondrial protein, known previously as Perm1, as a highly phosphorylated factor regulated during heart development. We renamed Perm1 as MICOS complex subunit Mic85 since it shows robust physical interaction with MICOS complex subunits, including Mitofilin (Mic60), Chchd3 (Mic19), Chchd6 (Mic25) and the outer membrane protein Samm50. Moreover, Mic85 is localized to the mitochondrial inner membrane facing the intermembrane space and the dynamics of Mic85 protein expression is regulated by the ubiquitin-proteasomal system through phosphorylation of casein kinase 2 on its PEST motif. Silencing of Mic85 in cultured neonatal cardiomyocytes impairs mitochondrial morphology and compromises oxidative capacity. Our findings support a clear role for Mic85 in the maintenance of mitochondrial architecture and in its contribution to enhanced energetics during developing and adult mouse cardiomyocytes. The transgenic Mic85 knockout mouse generated with a GFP knock-in will support future in vivo investigations on the integrity of mitochondria and the function of Mic85 in cardiac development.
Municipal wastewater contains nutrients valuable for a reuse in agriculture and can be the source of a multitude of chemicals used in private households and industry, too. As many of these chemicals are incompletely degraded during wastewater treatment, their residues remain partly in sewage sludge and partly in treated wastewater. Concerns are linked particularly to the so called micropollutants, i.e. anthropogenic organic substances such as personal care products, pharmaceuticals and biocides, for which scarce data on their degradability and environmental fate and particularly on their ecotoxicity are available. Thus, when reusing treated wastewater and sewage sludge for irrigation or as soil amendment for a sustainable land and water management, these wastewater-borne pollutants may enter soil, groundwater and surface water. The present work therefore aimed at assessing potential ecotoxic effects on aquatic and terrestrial organisms of reusing treated wastewater and sewage sludge. To this end, established as well as newly developed experimental approaches were used to investigate the problem on several levels. Individual wastewater-borne substances, samples from field study sites and samples from a soil column experiment simulating prolonged wastewater irrigation were examined.
At the start of the experimental work, the ecotoxicity of climbazole was characterised towards five aquatic and five terrestrial test organisms. Climbazole is an azole antimycotic agent applied in cosmetics and anti-dandruff shampoos and was recently detected in relatively high concentrations in treated wastewater and sewage sludge. In the present work climbazole was found to be particularly toxic towards plants such as water lentils with effective concentrations comparable to those of agricultural azole fungicides. Dwarfism, that is reduced shoot elongation observed in plants, pointed at a specific, phytohormone inhibiting mode of action of climbazole. Furthermore, the expected influence of the soil pH on the phytotoxicity of climbazole was experimentally confirmed.
Based on the findings for climbazole, two additional azole antimycotics, ketoconazole and fluconazole, and the regularly in sewage sludge detected biocide benzyldimethyldodecyl-ammonium chloride (BDDA) were investigated for their toxicity towards plants.
In aqueous medium, an increasing phytotoxicity from fluconazole to BDDA, ketoconazole and climbazole was observed, while in soil, phytotoxicity increased from BDDA to ketoconazole, climbazole and fluconazole. The relatively low terrestrial toxicity of BDDA and ketoconazole probably resulted from their strong binding to soil as well as their good biodegradability. To render the exposure scenario more realistic, sewage sludge was co-applied with the four test substances in a parallel test run. However, as no detectable influence on their effective concentrations was found, it can be assumed that the current practice of assessing sewage sludge borne substances with biotests in standard soil is sufficiently realistic. In a further study, different advanced sludge-treatment technologies were assessed for their efficacy in reducing pollutants. Results from the present work indicated that effects assessed in terrestrial short term biotests only seldom correlated with the concentrations of certain pollutants. Rather, a negative correlation of the stability of the sludges, determined by the ratio of volatile to total solids, to their ecotoxicity was seen.
Another aspect of the present work was the design and performance of an experimental approach to assess the environmental risk of a long-term irrigation with treated wastewater concerning the quality of soil and water in a prospective way, i.e. before the installation at field scale. For the simulation of a continuous irrigation corresponding to approximately 30 years, a percolation apparatus was developed and four different soils were percolated with treated wastewater for three months. Acute and chronic biotests with nine test organisms from different trophic levels (green algae, water lentils and water fleas as well as oilseed rape, oats, bacteria, spring tails, enchytraeids and earthworms) were used to assess the soil percolates as well as the soils with and without percolation. These investigations were accompanied by a comprehensive chemical monitoring conducted by project partners. Results indicated that the soil passage, that is the percolation through the soil, generally improved the quality of the treated wastewater as habitat for aquatic organisms which was visible by a reduction of its phytotoxicity. However, in some cases it deteriorated the water quality, probably resulting from the leaching of metals from pre-contaminated soil. A deteriorated habitat quality of the soil after the percolation with treated wastewater was observed for several test organisms and soils. In the same, mainly peaty soils, the highest accumulation of wastewater-borne micropollutants and of zinc was measured. Yet, their concentrations did not correlate to the observed biological effects. Moreover, data on ecotoxicity were only available for a small fraction of the detected substances so that their concentrations could not successfully be used to predict expected biological effects.
The experimental approach used in the present work demonstrated to be an adequate tool to support the prospective evaluation of environmental risks of treated wastewater irrigation. Overall, it can be concluded that the reuse of treated wastewater on soil can improve the quality of treated wastewater but that this can come at the cost of deteriorating the quality of the soil. As these risks cannot be generalised, a comprehensive biotest battery as well as chemical analysis should be used to assess them on a case-specific basis for each respective wastewater and the respective soil.
Seit mehr als 200 Jahren gibt es durch die Wissenschaftler der Wetterstation auf dem Hohenpeißenberg systematische Wetterbeobachtungen, doch erst seit wenigen Jahren gibt es unter den Klimaforschern einen Konsens, dass sich das Klima durch anthropogene Einflüsse schon verändert hat – und weiter verändern wird. Die bisherigen Auswirkungen, wie zum Beispiel ein globaler Temperaturanstieg von 0,85°C seit Beginn der Industrialisierung, sind heute gut belegbar. Mögliche zukünftige Entwicklungen des Klimas werden heute ebenso erforscht wie die Auswirkungen des Klimawandels auf Mensch und Umwelt. Zu diesen Auswirkungen gehören unter anderem Folgen für die Landwirtschaft. Durch veränderte Niederschläge und den Temperaturanstieg werden sich die Lebensbedingungen von Bodenorganismen und Anbaubedingungen für Pflanzen ändern. Letztendlich ist aufgrund dieser Veränderungen auch ein verstärkter Einsatz von Pestiziden zu erwarten. Allerdings wurde bisher kaum untersucht, ob der Einsatz von Pestiziden in der Landwirtschaft unter den Bedingungen des Klimawandels (konkret durch die Interaktion von klimatischen und chemischen Faktoren) ein erhöhtes Umweltrisiko für Bodenorganismen darstellt. Bisher werden klimatische Faktoren bei den Tests für die Zulassung von Pestiziden nicht berücksichtigt.
Daher wurde diese Fragestellung in der hier vorliegenden Dissertation am Beispiel der Effekte von zwei zugelassenen Pestiziden auf Bodenorganismen unter verschiedenen klimatischen Bedingungen untersucht. Konkret wurden dazu mit Labor- und Halbfreilandversuchen die Wirkung eines Insektizids und eines Fungizids in Interaktion von Temperatur und Bodenfeuchte auf Vertreter zweier Invertebratengruppen (Collembola: zwei Arten; Enchytraeidae: eine Art) untersucht.
In einem modifizierten Standardtest mit Collembolen erhöhte sich die Toxizität des Insektizids Lambda-Cyhalothrin, wenn die Exposition der beiden Arten bei einer erhöhten Bodenfeuchte stattfindet. Die kühl adaptierte Art Folsomia candida reagierte bei erhöhter Testtemperatur am empfindlichsten auf diese Testsubstanz: Die EC50 aus diesem Experiment lag bei 2,84 mg (a.s.)/kg Boden Trockengewicht (dw). Unter Standardbedingungen, wie sie in Tests für die Zulassung von Pestiziden angewandt werden, lag die EC50 von F. candida dagegen bei 8,65 mg a.s./kg dw.
Unter den gleichen Versuchsbedingungen wurde auch das Fungizid Pyrimethanil an Collembolen getestet. Hier erwies sich die Testsubstanz für beide Arten bei
gleichzeitigem Trockenstress und / oder erhöhter Temperatur als toxischer im Vergleich zu den Standard-Testbedingungen. Dabei zeigte F. candida mit einer EC50 von 28,3 mg a.s./kg dw die höchste Empfindlichkeit. Ohne die veränderten klimatischen Faktoren, betrug die EC50 von F. candida 52,3 mg a.s./kg dw.
In Reproduktionstests mit der Enchytraeen-Art Enchytraeus bigeminus wurde die Bodenfeuchte als klimatischer Faktor in Kombination mit jeweils einer Testsubstanz untersucht. Bei beiden Chemikalien reagierte E. bigeminus in trockenem Boden empfindlicher. Die ermittelten EC50 betrugen 1,34 mg a.s./kg dw für Lambda-Cyhalothrin und 437 mg a.s./kg dw für Pyrimethanil. Getestet unter Standardbedingungen lagen die EC50-Werte bei 3,79 bzw. 499 mg a.s./kg dw.
Neben den Laborexperimenten wurden Tests in „Terrestrischen Modellökosystemen“ (TME) mit den gleichen Chemikalien in Kombination mit variierender Bodenfeuchte als klimatischer Faktor vorgenommen. Diese Experimente wurden in Deutschland und in Portugal durchgeführt, um die Reaktion einer zentraleuropäischen und einer mediterranen Artengemeinschaft zu untersuchen. Aus der terrestrischen Lebensgemeinschaft wurden verschiedene Organismengruppen untersucht. Die Effekte auf Enchytraeen aus dem Experiment mit Pyrimethanil waren als Veröffentlichung Teil dieser Dissertation. In der portugiesischen Halbfreilandstudie wurden keine Effekte auf die Enchytraeen durch Pyrimethanil bei umweltrelevanten Konzentrationen festgestellt, jedoch beeinflusste die Bodenfeuchte die Zusammensetzung der Artengemeinschaft. Im deutschen TME-Experiment wurde eine verstärkte Wirkung des Fungizids in trockenem Boden festgestellt, d.h. die jeweiligen Effektkonzentrationen (niedrigste EC50 3,48 mg a.s./kg dw für Fridericia connata in trockenem Boden) lagen deutlich unterhalb der aus den Labortests mit Enchytraeus bigeminus bekannten Werten (499 mg a.s./kg dw).
Zusammenfassend lässt sich feststellen, dass klimatische Faktoren die Effekte von Pflanzenschutzmittel auf Bodenorganismen beeinflussen können. Für Laborversuche ist eine generelle Berücksichtigung von klimatischen Faktoren im Zulassungsverfahren aus heutiger Sicht zu weit gegriffen. Die TME-Versuche zeigten sich als geeignetes Testverfahren, interaktive Effekte von Pestiziden und Klima bzw. multiplen Stressoren generell auf Artengemeinschaften zu untersuchen. Für TME-Experimente wäre unter Beachtung der Vielzahl möglicher Fragestellungen, Endpunkte und moderner statistischer Auswerteverfahren eine internationale Richtlinie wünschenswert.
Biodiversity is unevenly distributed on Earth. Highly diverse biotas are particularly expected in mountain systems, because altitudinal zonation provides a number of habitat alternatives, which could lead to lower extinction rates during climatic changes. Nevertheless, the impact of environmental changes on plant diversification (especially for sub-alpine taxa) in the course of mountain orogenesis remains poorly understood. This is also true for the highest and largest plateau on Earth, the Qinghai-Tibetan Plateau (QTP) and its surrounding areas.In this doctoral thesis, I investigated the impact of environmental changes on plant diversification and the floristic exchange between the QTP region and biodiversity hotspots of Southeast Asia as well as other parts of the world by using the sub-alpine genera Agapetes and Vaccinium (Vaccinieae, Ericaceae) as well as Tripterospermum (Gentianinae, Gentianaceae) as model systems. Furthermore, I examined the role of niche evolution and conservatism in a changing environment over time, and detected possible beneficial morphological traits of plants in the surroundings of the QTP by investigating subtropical Gentianinae (Crawfurdia, Kuepferia, Metagentiana, Sinogentiana, and Tripterospermum; Gentianaceae).
Effekte von Neonikotinoiden auf die Aktivität des Muskels M17 und das Lernverhalten der Honigbiene

Im Rahmen dieser Arbeit wurden Untersuchungen zur Auswirkung von Neonikotinoiden auf die Muskelspikeaktivität des Muskels M17 und auf das Lernvermögen in einer komplexen Aufgabe an der Honigbiene (Apis mellifera carnica) durchgeführt. Dabei wurden drei verschiedene Substanzen verwendet: Clothianidin, Thiacloprid und Imidacloprid. Neonikotinoide stehen häufig im Verdacht, für das Sterben von Bienenvölkern verantwortlich zu sein, da die Bienen bei ihrer Nahrungssuche an behandelten Pflanzen den Substanzen ausgesetzt sind und diese möglicherweise damit auch an ihr Volk weitergeben. Die vorliegende Arbeit soll dazu beitragen, diese mögliche Gefährdung der Honigbiene durch Neonikotinoide weiter aufzuklären und deren Risiken zu beurteilen. Der Hintergrund für die Versuche zur Muskelspikeaktivität waren vorangegangene Versuche, die Auswirkungen von Neonikotinoiden auf die Motorik von sich frei bewegenden Individuen dokumentierten. In der vorliegenden Arbeit wurde untersucht, ob sich diese Effekte auch an der Spikeaktivität des Muskels M17 widerspiegeln. Dafür wurden Elektromyogramme des Muskels M17 zu verschiedenen Zeitpunkten nach der Gabe der Substanzen erstellt und deren mediane Anzahl mit einer Kontrollgruppe verglichen.
In einem ersten Versuch wurden Clothianidin (1 µM), Thiacloprid (1 µM), Imidacloprid (1 µM) oder eine Kontrollsubstanz (PBS) in die Kopfkapsel appliziert. Beim Vergleich mit der Kontrollgruppe zeigten sich für Imidacloprid keine Auswirkungen. Clothianidin verursachte eine deutlich erhöhte mediane Spikerate des Muskels M17, während Thiacloprid diese absenkte, beides im Vergleich zur Kontrollgruppe. Auch bei einer um 30 Minuten versetzten Doppelapplikation von Clothianidin (1 µM) und Thiacloprid (10 µM) stellten sich diese Effekte ein, wobei Clothianidin eine dominante Rolle einzunehmen scheint. Diese Ergebnisse stehen im Einklang mit Untersuchungen zur Laufaktivität, welche sich ebenfalls durch Clothianidin erhöhte und durch Thiacloprid absenkte.
Ein weiteres Experiment untersuchte die Auswirkung einer akuten Fütterung mit Clothianidin (1 ng in 1 µl) bzw. Thiacloprid (250 ng in 1 µl) auf die Anzahl der Muskelaktionspotenziale. Auch hier zeigt sich eine deutliche Erhöhung der Spikeanzahl durch Clothianidin und eine Absenkung der Spikeanzahl durch Thiacloprid, was die Ergebnisse des ersten Versuchs nochmals bestätigt.
Des Weiteren wurden Untersuchungen zur Auswirkung einer chronischen Fütterung mit Clothianidin (50 ppb) bzw. Thiacloprid (5000 ppb) auf die Anzahl an Spikes durchgeführt. Die Bienen wurden dabei über mehrere Wochen im Volk mit den jeweiligen Substanzen gefüttert. Dabei zeigte sich, dass auch eine chronische Fütterung der Bienen mit Clothianidin ihre Anzahl an Muskelaktionspotenzialen deutlich erhöht, während eine chronische Fütterung mit Thiacloprid diese absenkt. In einer Kombination der chronischen Fütterung mit einer zusätzlichen akuten Fütterung des jeweils anderen Neonikotinoids zeigte sich, dass auch hier Clothianidin eine dominante Rolle gegenüber Thiacloprid einnimmt und auch keine synergistischen oder Gewöhnungseffekte der beiden Substanzen eintreten. Die chronisch eingefütterten Völker entwickelten sich dagegen wie die Kontrollvölker, sodass die hier beschriebenen Auswirkungen auf die Einzelbiene keine sichtbaren Effekte auf ganze Völker zu haben scheinen.
Zusätzlich wurden Versuche zum Duftlernen in einer komplexen Lernaufgabe, dem positiven Patterning, unter Einfluss von Clothianidin durchgeführt. Die Bienen müssen dabei zwischen unbelohnten Einzeldüften und einem belohnten Duftgemisch, bestehend aus den beiden Einzeldüften, unterscheiden. In verschiedenen Versuchsdurchläufen wurden 0,25 ng oder 1 ng Clothianidin (jeweils 1 µl) in den Thorax injiziert, entweder vor der Akquisitionsphase oder vor dem ersten Abruftest nach drei Stunden. In keinem der Versuchsdurchläufe zeigten sich Effekte des Clothianidins auf den Lernvorgang, weder in der Akquisitionsphase noch in den Abruftests. Somit wurden weder das Lernen an sich, noch die Konsolidierung und damit die Überführung des Gelernten in das Langzeitgedächtnis durch das Clothianidin beeinflusst. Allerdings könnte die Immunabwehr der Bienen nach längerer Einwirkung des Clothianidins (24 h) in der höheren Konzentration herabgesetzt sein, da viele Bienen starben.
Insgesamt ergaben sich Effekte von Clothianidin und Thiacloprid auf die Anzahl der Aktionspotenziale des Muskels M17, die sich aber im gesamten Volk nicht widerspiegeln. Der Lernvorgang in der hier durchgeführten komplexen Lernaufgabe wird durch Clothianidin nicht beeinflusst. Möglicherweise entsteht dieser Unterschied in den Ergebnissen durch eine Bindung der Substanzen an verschiedene Rezeptorsubtypen, die pharmakologisch unterschiedliche Eigenschaften besitzen und somit auch unterschiedliche Auswirkungen zeigen.
Im Rahmen der hier vorliegenden Arbeit wurde der Einfluss sowohl akuter als auch chronischer Aufnahme subletaler Mengen des Insektizids Thiacloprid auf Einzelbienen und Bienenvölker untersucht. Anlass für diese Art an Untersuchungen gibt ein seit Jahren in Nordamerika und Europa auftretendes unerklärliches Phänomen, „Colony Collapse Disorder“ genannt, bei dem Bienenvölker durch einen plötzlichen Verlust der Flugbienen zusammenbrechen. Als Ursache für das Völkersterben stehen neben anderen Faktoren wie Parasiten, Pathogenen und Umweltfaktoren die Insektizide aus der Gruppe der Neonikotinoide und deren Auswirkungen auf Bienen in subletalen Mengen im Verdacht. Basierend auf Studien der Europäischen Behörde für Lebensmittelsicherheit (EFSA) wurde der zugelassene Einsatz der drei Neonikotinoide Clothianidin, Imidacloprid und Thiamethoxam im Pflanzenschutz für zunächst zwei Jahre durch die EU-Kommission stark eingeschränkt.
Thiacloprid, ein weiteres Insektizid, welches zur Gruppe der Neonikotinoide gehört, ist weiterhin für den Einsatz im Pflanzenschutz zugelassen. Es wirkt in ähnlicher Weise wie die zuvor genannten Neonikotinoide als Agonist am nikotinischen Acetylcholinrezeptor, wobei es jedoch als weniger toxisch für Bienen gilt. Trotzdem sind subletale Auswirkungen dieses Neonikotinoids auf Bienen denkbar, die sich in Verhaltensänderungen der Bienen äußern und als Folge Einfluss auf das gesamte Bienenvolk nehmen könnten.
In der hier vorliegenden Arbeit wurden in chronisch mit Thiacloprid eingefütterten Völkern über mehrere Monate regelmäßige Populationsschätzungen durchgeführt, um die Entwicklung der Bienenvölker unter Aufnahme von Thiacloprid festzustellen. In einem weiteren Versuch wurde die Entwicklung der Brut unter chronischer Fütterung mit Thiacloprid beobachtet. Zusätzlich wurde eine große Zahl an Bienen mit RFID-Transpondern ausgestattet, um das Flugverhalten zu dokumentieren. Insbesondere wurden hier der Zeitpunkt des ersten Ausflugs und die Lebensdauer der Bienen zu Vergleichen herangezogen. Nach akuter Fütterung einer subletalen Einzeldosis Thiacloprid wurden Versuche zum Heimkehrvermögen von Bienen durchgeführt.
Unter feldrelevanten und bis zu zehnfach höheren Thiacloprid-Konzentrationen wurden keine beeinträchtigenden Einflüsse auf die Volksentwicklung beobachtet. Bei Konzentrationen, die um ein 25faches bzw. ein 40faches höher als die feldrelevante Konzentration waren, wurde festgestellt, dass die Brutzellenanzahl im Verhältnis zur Bienenanzahl verringert war. Bienen aus chronisch mit Thiacloprid eingefütterten Völkern starteten mit höherem Alter zu ihrem ersten Flug aus dem Bienenstock. Die Zeit, die die Bienen als Sammlerinnen verbrachten, änderte sich nicht. Durch Beobachtungen der Brutflächen konnte festgestellt werden, dass sich die Brut in Thiacloprid-gefütterten Völkern entsprechend der Brut in Kontrollvölkern entwickelte. Aufgrund weiterer Ergebnisse wurde eine Störung der olfaktorischen Wahrnehmung von Bienen aus Thiacloprid-gefütterten Völkern vermutet. Die akut verabreichte subletale Dosis an Thiacloprid führte zu einem erheblichen Verlust an heimkehrenden Bienen und deutet auf eine Beeinträchtigung des Orientierungs- bzw. Navigationsvermögens der Bienen hin.
In den durchgeführten Versuchen wurden sowohl direkte Auswirkungen von chronischer und akuter Aufnahme subletaler Mengen an Thiacloprid, als auch indirekte Auswirkungen auf Honigbienen beobachtet. Da teilweise erst bei hohen, nicht feldrelevanten Konzentrationen in den Versuchen Effekte beobachtet wurden, kann nur bedingt durch die Verhaltensänderung von Einzelbienen auf daraus resultierende Auswirkungen auf ein gesamtes Bienenvolk unter realistischen Bedingungen geschlossen werden.
Rotary adenosine triphosphate (ATP)ases are ubiquitous, membrane-bound enzyme complexes involved in biological energy conversion. The first subtype, the so-called F1Fo ATP synthase, predominantly functions as an ATP synthesizing machinery in most bacteria, mitochondria and chloroplasts. The vacuolar subtype of enzyme, the V1Vo ATPase, operates as an ATP driven ion pump in eukaryotic membranes. The subtype found in archaea and some bacteria is called A1Ao ATP (synth)ase and is capable of working in both directions either to synthesize ATP or to generate an ion motive force by consuming the same.
All the three above-mentioned subtypes of rotary ATPases work as nanomolecular machines sharing a conserved mechanism to perform the energy conservation process. The simplest form of these enzymes is the bacterial F1Fo ATP synthase. Here, ions are channelled via the membrane stator subunit a to the rotor ring of the enzyme. After almost a complete rotation of the ring the ions are released again on the other side of the membrane. This rotation is further transmitted via the central stalk to the soluble part of the enzyme, the F1-complex, where conformational changes within the nucleotide binding sites result in the synthesis of ATP from ADP and Pi.
The rotor or c-ring of the enzyme is the key protein complex in mediating transmembrane ion translocation. Several structural and biochemical methods have been applied in the past years to study the rotor rings from many different organisms. The results revealed that the stoichiometry of a c-ring of a given species is constant while it can vary between different species within a range of 8 to 15 c subunits. The c-ring stoichiometry determines directly the number of ions transported through Fo per rotation whereby three molecules of ATP are concurrently synthesized in the water-soluble F1 headgroup. Hence the number of c subunits has an important influence on the bioenergetics of the corresponding enzyme and thus the entire organism.
The c-ring of a rotary ATPase is able to specifically bind either protons (H+) or sodium ions (Na+) as the coupling ion for the enzyme. Several structures are already available revealing the coordination network of both types of rotor rings. In each case ion binding includes a highly-conserved carboxylic acid residue (glutamate or aspartate), in addition to a more varying combination of amino acid residues, whereby Na+ coordination is structurally more demanding than H+ binding.
In the first part of my PhD thesis, I aimed to characterize the F1Fo ATP synthase rotor ring of the opportunistic pathogenic bacterium Fusobacterium nucleatum on a functional and structural level. F. nucleatum is an anaerobic bacterium which uses peptides and amino acids as a primary energy source. It is one of the most frequently occuring bacteria in human body infections and involved in human periodontal diseases.
The protein complex was heterologously expressed within a hybrid ATP synthase in Escherichia coli and purified without an affinity tag for further analysis. Two high resolution X-ray structures of the c-ring were solved at low (5.3) and high (8.7) pH to 2.2 and 2.64 Å, respectively. In both structures, the conserved glutamate is in an ion-locked conformation, revealing that the conformational state of the ion binding carboxylate is not depending on the pH of the crystallization condition, which is in good agreement with previous structural and biochemical studies of other c-rings.
A Na+ ion is present within the c-ring binding site and directly coordinated by four amino acid residues and a structural water molecule. Remarkably, the Na+ is bound by two glutamate residues instead of one as is the case in the I. tartaricus Na+ binding c-ring, of which the first high resolution X-ray structure of a c-ring has been solved in 2005. Thus, a new type of Na+ coordination in an ATP synthase rotor ring with a two-carboxylate ion binding motif is described here, which also occurs in other bacteria, including several pathogens. Na+ specificity of the investigated c-ring was further confirmed by a competitive biochemical labeling reaction performed with a fluorescent ATP synthase inhibitor molecule (N-cyclohexyl-N`-[4(dimethylamino)-α-naphtyl] carbodiimide, NCD-4).
We furthermore complemented our functional and structural data of the F. nucleatum c-ring by computational studies to explore the ion translocation mechanism of this enzyme in more details. We therefore analyzed the protonation state of the second, additional glutamate in the ion binding site. Molecular dynamics (MD) simulations and free-energy calculations indicated that this glutamate is constitutively protonated, in the ion-locked as well as in a simulated, more hydrated open-conformation of the ion binding glutamate as when it is travelling through the a/c-ring interface upon c-ring rotation.
Resistance in glucocorticoid-induced apoptosis is associated with poor prognosis for long term survival in childhood acute lymphoblastic leukemia (ALL). As Smac mimetics have been shown to reactivate apoptosis by antagonizing Inhibitor of Apoptosis (IAP) proteins, we investigate the potential of the Smac mimetic BV6 to overcome glucocorticoid-resistance in ALL. This study shows that BV6 synergistically cooperates with glucocorticoids to trigger apoptosis and to suppress clonogenic growth of pediatric ALL cells. Of note, the BV6/glucocorticoid combination treatment also induces cell death in cells having defects in the apoptotic signaling cascade by inducing a switch from apoptotic to necroptotic cell death. The clinical relevance of our novel combination treatment is underscored by parallel experiments in primary pediatric ALL samples, in which glucocorticoids and BV6 act together to induce cell death in a synergistic manner. Importantly, the addition of BV6 enhances the anti-leukemic effects of glucocorticoids in an in vivo mouse model of pediatric ALL without causing substantial side effects, highlighting the potency of a BV6/glucocorticoid combination treatment. In contrast, BV6 does not increase cytotoxicity of glucocorticoids against several non-malignant cell types of the lympho-hematopoietic system. Furthermore, we have identified the novel underlying mechanism of BV6/glucocorticoid-induced apoptosis by showing that BV6 and glucocorticoids synergistically act together to promote assembly of the ripoptosome, a RIP1/FADD/caspase-8-containing cell death complex. Ripoptosome assembly is critically required for BV6/Dexamethasone-induced cell death, since genetic silencing of its members, i.e. RIP1, reduces ROS production, caspase activation and most importantly cell death induction. BV6/glucocorticoid combination treatment promotes ripoptosome assembly by inhibition of both of its negative regulators, IAP proteins and cFLIP. Thus, we identify that BV6 and glucocorticoids cooperate together to reduce cIAP1, cIAP2 and XIAP protein levels and cFLIP expression. Ripoptosome formation occurs independently of autocrine/paracrine loops of death receptor ligands, since blocking antibodies for TNFα, TRAIL or CD95L or genetic silencing of their corresponding receptors fail to rescue BV6/glucocorticoid-induced cell death. In summary, this study shows that the Smac mimetic BV6 sensitizes for glucocorticoid-induced apoptosis by promoting ripoptosome assembly with important implications for the treatment of childhood ALL.
Die hier vorliegende Dissertation befasst sich mit der Synthese von Naturstoffen aus Xenorhabdus und Photorhabdus spp. Da 6,0 - 7,5% ihres Genoms Sekundärmetabolit Clustern zuzuordnen sind, gelten diese entomopathogenen Bakterien als vielversprechende Naturstoffproduzenten. Die Palette der von ihnen produzierten Naturstoffe reicht von Antibiotika über Insektizide bis hin zu potentiellen Zytostatika. Die im Rahmen dieser Arbeit synthetisierten und charakterisierten Substanzen lassen sich in vier Kategorien einteilen: kleine Sekundärmetabolite (Phurealipide), zyklische Makrolaktame (Xenotetrapeptide, GameXPeptide und Ambactin), zyklische Makrolaktone (Szentiamide, Xentrivalpeptide und Xenephematide) und methylierte lineare Peptide (Rhabdopeptide und Rhabdopeptid-ähnliche Moleküle).
The timing and duration of leaf deployment strongly regulate earth-atmosphere interactions and biotic processes. Leaf dynamics therefore have major implications for life on earth, including the global energy balance, carbon and water cycles, feedbacks to climate, species extinction risk and agriculture. Evidence of shifts in the timing of leaf deployment and senescence (leaf phenology) as a result of climate change has been accumulating over the past decades, particularly in relation to spring phenology in the northern hemisphere. However, leaf phenological change in other parts of the world has received less attention. This thesis quantifies global phenological change over the past three decades using remotely sensed data. Phenological change was found to be widespread and severe, also in the southern hemisphere. While the detected change testifies of the phenological plasticity of many plant species, it is not clear if the duration of leaf deployment (leaf habit) is equally sensitive to environmental change. Since evergreen and deciduous leaf habits are often distinctly sorted along environmental gradients, ecologists have hypothesised that these patterns result from natural selection for an optimal leaf habit, under a given environmental regime. Such evolutionary convergence can be examined by testing if the physiological niche that is occupied by a particular leaf habit (evergreen or deciduous) is similar among regions with distinct evolutionary histories. Using a process-based model of plant growth and a constructed map of evergreen and deciduous vegetation, the physiological niche of leaf habits was quantified in four global biogeographic realms. Substantial niche overlap was found between the same leaf habit in different realms, suggesting evolutionary convergence of the physiological niche. This implies a sensitivity of leaf habit to environmental change, as environmental variables determine the geographic space where the physiological niche allows a positive carbon balance, and therefore occurrence of the leaf habit. Since the physiological niche consists of the integrated effects of physiological traits and trade-offs, environmental dependencies and leaf habit and phenology, an understanding of the carbon economy of individual plants requires decomposing the physiological niche into its components. Using empirical data on leaf phenology, leaf habit and physiological processes from woody species in a seasonally dry African savanna, a simple carbon balance model was parametrised. Carbon gain varied considerably between species as a result of substantial variation in leaf habit, leaf phenology and physiological traits. The multiple lines of evidence in this thesis therefore suggest that, while convergent selective forces may determine the dominant leaf habit in a particular environment, inter-specific variation is substantial, potentially as a consequence of historical contingencies or competitive interactions.
Habituation ist eine der einfachsten Formen des Gedächtnisses. Hierbei handelt es sich um die erlerne Gewöhnung an einen harmlosen Reiz. Dies bedeutet, dass nach mehrfacher wiederholter Repräsentation eines harmlosen Reizes die Reaktion darauf stetig abnimmt, bis sie völlig zum erliegen kommt. Je nach Trainingsprotokoll kann diese Gewöhnung bis zu mehren Tagen andauern. Habituation ist hoch konserviert und ein Verhaltensmuster, dass auch bei sehr einfachen vielzelligen Organismen zu finden ist und untersucht werden kann. Zur Untersuchung des Zusammenspiels innerhalb eines neuronalen Netzwerkes, welches für die Habituation des Rückzugsreflexes (Ausweichreaktion nach Berührung) verantwortlich ist wurde hier der Fadenwurm Caenohabditis elegans (C. elegans) als Modell Organismus verwendet. Aufgrund seines einfachen, nur 302 Zellen umfassenden, Nervensystems eignet sich C. elegans sehr gut für Grundlagenforschung in diesem Bereich. Das neuronale Netzwerk, das verantwortlich ist für den Rückzugsreflex ist in drei Ebenen organisiert. Wahrgenommen wird der Reiz von sensorischen Neuronen (ASH, ALM, AVM, PLM, PVM). Die Weiterleitung erfolgt über verschiedene Interneuronen (AVA, AVB, AD, AVE, PVC) hin zu den Motorneuronen, welche die Muskeln enervieren und somit die Reaktion auf den in erster Ebenen wahrgenommen Reiz auslösen.
Mit Hilfe von optogenetischen Werkzeugen wurde hier Untersucht welche Rolle einzelne Zellen innerhalb dieses Netzwerkes innehaben und an welcher Stelle innerhalb des Netzwerkes die kurzzeitige Habituation des Reizes, nach einem Einfachen Lernprotokoll stattfindet. Zuerst musste eine Möglichkeit gefunden werden die zur Verfügung stehenden optogenetischen Werkzeuge zellspezifisch zu exprimieren. In dieser Arbeit wurden hierfür Rekombinasesysteme verwendet, die es ermöglichten zur Expression eine Kombination aus 2 verschiedenen Promotoren zu verwenden. Beide Promotoren dürfen hierbei nur in einer Zelle, der Zielzelle, überlappen. Es konnte zellspezifische Expression des Kationenkanals Chanelrhodopsin 2 (ChR2) in den beiden Zellparen AVAL/R und ASHL/R (nimmt aversive Reize wahr) erreicht werden.
Zur Untersuchung der Habituation wurde zusätzlich noch ein Wurmstamm verwendet, welcher ChR2 unter dem mec-4 Promotor exprimiert. ChR2 ist hier in den Mechanorezeptorneuronen (MRN) ALM, AVM, PLM und PVM exprimiert. Die hier durchgeführten Experimente deuten darauf hin das den MRNs die Größte Rolle bei der Ausbildung einer Habituation zukommt. Es gibt jedoch auch Hinweise darauf, dass AVA zusätzlich eine Rolle spielt.
Im weiteren Verlauf der Arbeit wurde die Rolle von AVA genauer untersucht. AVA gilt als der Hauptsignalgeber für eine Rückwärtsbewegung (spontan und nach Reizempfang). Es konnte gezeigt werden dass eine Unterbrechung der ’Gap Junktionen’ zwischen AVA und PVC eine stärkere Reaktion zur Folge haben. AVA scheint also durch PVC inhibiert zu werden. Ebenfalls mit AVA direkt interagierende Neuronen sind AVD und AVE. Mit den hier zur Verfügung stehenden Mitteln konnte die genaue Modulation von AVA durch diese Zellen jedoch nicht gezeigt werden.
In dieser Arbeit konnte der Grundstein für eine funktionale Aufklärung des Nervensystems von C. elegans gelegt werden. Vor allem durch die Möglichkeit der zellspezifischen Expression kann es zukünftig gelingen das Zusammenspiel der einzelnen Nervenzellen und ihren Anteil an einem bestimmtem Verhalten zu Untersuchen.
The mammalian family of bears (Ursidae) comprises eight extant species, occurring on four different continents. Among them are the iconic and well-known brown and polar bears, both widely distributed across the Northern hemisphere. Their intraspecific genetic structuring has been extensively investigated, albeit with a focus on genetic markers from maternally inherited parts of their genomes (mitochondrial DNA). The evolutionary relationship and divergence time between brown and polar bears have recently triggered an extensive debate, while less focus has been put on to other parts of the ursid phylogeny, particularly to a clade of three Asian bear species. To date, whole genomes of more than 100 bear individuals from four different species have been sequenced. Yet, one fundamental part of the genome has been largely omitted from specific analyses, in bears as well as in most other mammals: the Y chromosome.
The mammalian Y chromosome provides a unique perspective on the evolutionary history of organisms due to its distinct features, and specifically reflects the patriline because of its male-specific inheritance. The characteristics of this chromosome make it well suited to complement and contrast evolutionary inferences based on other genetic markers, and to uncover processes like sex-biased gene flow and hybridization. The unique insights that can be gained from analyses of Y-linked genetic variation made me utilize this part of the genome to investigate the evolution of male lineages in bears. Studying the patriline is particularly promising in this taxonomic group because of male-biased dispersal and a complex and fast radiation of bears. The analysis of Y-chromosomal genetic markers is thus the common theme of this dissertation: I present the identification of large amounts of Y-chromosomal sequence, the development of male-specific markers from such sequences, and the application of these markers to trace the evolution of male lineages of different bear species.
Specifically, I developed a molecular sex determination system based on the detection of two Y-linked fragments that allows to reliably discriminate between females and males from seven different bear species (Bidon et al. 2013). The approach is highly sensitive, bear-specific, and can be applied in standard molecular laboratories. This makes it valuable in conservation genetics and forensic applications, e.g. to analyze non-invasively collected samples.
Furthermore, I used Y-linked markers in a comprehensive and range-wide sample of brown and polar bears, and show that male-biased gene flow plays an important role in distributing genetic material throughout the ranges of both species (Bidon et al. 2014). In brown bears, I detected a lack of paternal population structuring which is in strong contrast to the detailed structuring of the matriline.
Analyzing Y-chromosomal sequences from all eight bear species, I present a phylogeny of the patriline that largely resembles the topology from other nuclear markers but is different from the topology of the mitochondrial gene tree (Kutschera et al. 2014). This discordance among loci generates interesting hypotheses about inter-species gene flow, particularly among American and Asiatic black bears.
With the identification of almost two million basepairs of Y-chromosomal sequence and the analysis of an unprecedented large male-specific dataset in polar bears, a high-resolution view on the distribution of their intraspecific variation was obtained (Bidon et al. 2015). In particular, two clades that are divergent but do not show pronounced phylogeographic structure were detected, confirming the great dispersal capacity of males of this high arctic species.
This dissertation thus represents a comprehensive investigation of Y-linked genetic variation on the intra- and interspecific level in a non-model organism. With my research, I contribute to an increased understanding of the complex evolutionary history of bears. In particular, I show that male-biased gene flow strongly influences the distribution of nuclear genetic variation, and that the contrast between phylogenies of differentially inherited markers can help to understand interspecific hybridization between closely related species. Moreover, my findings demonstrate the potential of Y-chromosomal markers to uncover unknown evolutionary patterns and processes. This applies not only to bears but to many species, even such that are generally well known and well described.

Kieselalgen können auf hocheffiziente Weise Energie aus dem Sonnenlicht gewinnen. So überleben sie selbst lange Dunkelphasen im Meer. Doch wie schützen sie sich vor zu viel Strahlung, wenn Wind und Strömung sie in seichtes Wasser oder an die Oberfläche treiben? Dahinter steckt ein cleverer Regulations-Mechanismus.
Der unscheinbare Fadenwurm "C. elegans" ist einer der ersten und bis heute wichtigsten Modellorganismen der Optogenetik. Zwei Frankfurter Arbeitsgruppen gelang es vor zehn Jahren erstmals, das Tier genetisch mit lichtaktivierbaren Ionenkanälen auszustatten und seine Bewegungen mit Licht zu steuern. Inzwischen studieren Forscher an dem durchsichtigen Wurm auch Prozesse, die für die medizinische Forschung bedeutsam sind – etwa die Entstehung und Behandlung genetisch bedingter Herz-Rhythmus-Störungen.
The metabolome of any live cell consists of several hundred, if not thousands of different molecules at any given moment, be it a relatively small bacterial cell or a whole multicellular organism. Although there are continuous attempts to differentiate between primary and secondary metabolites, the borders often blur in the eye of almost perfect interconvertability of all such matter. With chemistry and physics dominating this domain of biology it is an interdisciplinary endeavor to tackle the questions surrounding the workings of the metabolic pathways involved, searching for answers that ultimately help us to better understand life and find solutions to problems that affect us humans. One area of biochemistry that serves as a formidable example of the intertwined primary and secondary metabolic pathways are fatty acids, essential components of bacterial membranes, sources of energy and carbon but also important building blocks of several natural products. The second area to be mentioned is the metabolism of amino acids, the basic components of proteins and enzymes, which also serve as precursors to a diverse set of metabolites with many biological purposes.
This work focuses on these two areas of biochemistry, as several intermediates of their metabolism serve as building blocks for complex secondary metabolites whence many interesting and bioactive natural products are derived. The powerful and relatively novel tool of click-chemistry is employed to track azide-labeled precursors of primary and secondary metabolism in various bacterial strains to observe biochemistry at work and adds to the knowledge gained through other methods. The methods presented in this work serve the observation of fatty acid biosynthesis, degradation, modification and transport through direct ligation of azido fatty acids with cyclooctynes on one hand, leading to a revision of fatty acid transport in general. On the other hand a cleavable azide-reactive resin is devised to generally track the fate of azidated compounds through the myriads of metabolic pathways offered by entomopathogenic bacteria possessing a rich secondary metabolism. The resulting findings led to the identification of several antimicrobial peptides, amides and other compounds of which many had remained so far undetected in the strains that underwent investigation, underlining the worth of this method for future metabolomic research and beyond.
Termites are important ecosystem engineers of the savanna biome, with the large mounds of fungus-cultivating termites being sources of habitat heterogeneity and structural complexity in African savanna landscapes. Studies from different localities throughout Africa have shown that termite mounds have a strong influence of diversity and composition of plant communities. However, most research has been conducted only at the local scale, and integrating knowledge across Africa is hampered by different methodology of studies and differing environmental context. Little is known about the variation in vegetation composition on termite mounds compared to the surrounding savanna at the regional scale and at the landscape scale, and the main determinants of plant communities on mounds are yet to be ascertained.
This thesis aimes at better understanding the influence of termite mounds on vegetation compared to the surrounding savanna across spatial scales. Three research projects analyse vegetation data and soil data from paired mound and savanna plots in West Africa. The first project examines the influence of termite-induced heterogeneity on plant diversity and vegetation composition at a regional scale, following a bioclimatic gradient from the Sahel of Burkina Faso to the Sudanian vegetation zone in North Benin. The second Project analysed variation of vegetation on and off mounds at the landscape scale in Pendjari National Park, North Benin. The third is a monitoring study over the course of two years, exploring dynamics of juvenile woody plant communities on mounds and in the surrounding savanna at a local scale. The thesis thus provides the first comparative quantitative analysis across scales of mound and savanna vegetation and the drivers of the mound–savanna difference in vegetation.
Synthesizing across scales, its results confirm that termite mounds strongly contribute to savanna plant diversity, even though mounds are not generally more species rich than the surrounding savanna. Variation in mound vegetation is much higher along climatic and soil gradients than previously acknowledged. Mound vegetation differs from the surrounding savanna in the whole study area and in each sampled savanna type, with the strongest differences occurring at the most humid study sites. A large proportion of the differences between mound and savanna vegetation is explained by clay enrichment and related soil factors, such as cation concentrations. Plants on mounds thus benefit from favourable soil conditions, including higher fertility and higher water availability, which is also mirrored by the higher abundance and basal area of juvenile woody plants found on mounds. The variation in mound vegetation between study sites across scales results in part from local differences in soil composition and from climatic differences that influence the regional distribution of species. Different sets of characteristic mound species are identified in each project. Specific plant families and traits like succulency, lianescence, and adaptations to zoochory are found to be overrepresented in mound communities.
In addition to the findings in this thesis, remaining parts of the variation in mound vegetation between study sites could likely be explained by investigating further factors. Specifically, mound vegetation depends on habitat context, which includes available species pools, spatial distribution of mounds, biotic interactions with dispersers and herbivores, fire, and also anthropogenic influence. The high proportion of species with adaptations to zoochory found on mounds, for example, indicates that animal dispersers should be of particular importance for vegetation on termite mounds. Herbivory and fire regime, which are known to contribute to the diversity and community composition of the mound–savanna system, also show strong local variation, not least because of anthropogenic influence.
In conclusion, termite mounds play a crucial role in maintaining heterogeneity and plant diversity in the savanna across scales. Ecosystem services provided by termites, especially considering long-term effects on soil fertility and ecosystem resilience, are most likely undervalued. Mounds should be considered in management plans from local to regional, transnational scales as a matter of course, accompanied by further research on the role of termite mounds in savanna ecology on a longer temporal scale. The research presented here thus provides a basis for future studies on termite mound vegetation that should specifically consider the biotic and abiotic context of the mound–savanna system.
Background. There is growing public and scientific concern about the occurrence of anthropogenic chemicals in the aquatic environment. Surface and groundwater serve as main drinking water resource. Especially in metropolitan areas these water reservoirs are impacted by organic pollutants predominantly originating from wastewater treatment plant (WWTP) effluents. The impact of wastewater derived anthropogenic chemicals is therefore related to environmental and human health concerns. In order to lower the potential environmental and human health risk from wastewater associated pollutants, strategies for enhanced pollutant removal are applicable in a medium-term perspective. Ozonation and powdered activated carbon treatment are the two advanced wastewater treatment technologies, which are technically mature as well as economically feasible for the application in large-scale wastewater treatment plants. While powdered activated carbon removes substances by adsorption, ozonation degrades a parent compound into oxidation products. Most of the available research has been done at lab-scale while onsite ecotoxicity tests and chemical analyses are rare.
Objectives. For a comparative evaluation of advanced wastewater treatments' potential to alter toxicity, a broad spectrum of ecotoxicological data need to be collected. The focus has been set on three major objectives: A) Evaluation of the endocrine activity; B) Evaluation of the unspecific toxicity; C) Evaluation of genotoxicity and mutagenicity.
Methods. The advanced treatment methods, ozonation and powdered activated carbon treatment of secondary wastewater effluents, – each equipped with subsequent sand filtration as additional post treatment step – were ecotoxico-logically characterized at a pilot-scale WWTP. For process control the elimination of 35 selected pharmaceuticals was identified by chemical analyses using HPLC-MS/MS.
The endocrine activity ((anti-)estrogenic, (anti-)androgenic, dioxin-like activity)) was characterized by yeast-based in vitro bioassays and cytotoxicity by cell based assays. Genotoxicity and mutagenicity was assessed using umuC'assay and Ames assay, respectively. All in vitro assays were performed using extracts of the wastewater samples. In vivo toxicity was assessed with the fish early life stage test with rainbow trout (Oncorhynchus mykiss). Ozonation was additionally assessed at a full-scale WWTP with in-vitro tests on endocrine activity and cytotoxicity and in vivo toxicity tests using five aquatic model organisms: Lemna minor, Daphnia magna, Chironomus riparius, Lumbriculus variegatus, Potamopyrgus antipodarum.
Results. In conventional activated sludge treated effluents the residual estrogenicity, antiandrogenicity, aryl hydrocarbon receptor agonistic activity and cytotoxicity were considerably reduced while antiestrogenicity was increased by both advanced treatment technologies. Ozonation led to an increase in genotoxic effects detected with Ames assay and with single cell gel electrophoresis of rainbow trout erythrocytes. Furthermore, mortality of rainbow trout was increased and reproduction of L. variegatus was decreased. Sand filtration lessened the genotoxic effects and adjusted reproduction of L. variegatus and mortality of rainbow trout to a similar level as conventional treatment.
Conclusions. This work demonstrates that conventional activated sludge treatment induces in vitro and in vivo toxicity. Advanced wastewater treatment combined with subsequent sand filtration can reduce in vitro and in vivo toxicity. An observed increase of endocrine activity after advanced wastewater treatment is an indication for different removal efficiencies of chemicals causing agonistic or antagonistic activity, respectively. Ozonation of wastewater generates ecotoxicity, which is largely removed by subsequent sand filtration. After a comprehensive investigation and after assurance of the removal of adverse effects, advanced treatment technologies could have beneficial effects on the ecological quality of the receiving water.
Die Endometriose ist eine gynäkologische Erkrankung, bei der epitheliale und stromale Zellen des Endometriums Läsionen außerhalb des Uterus bilden, die in ihrem Aufbau dem Endometrium gleichen. Diese Läsionen, sowie deren zyklische Proliferation, führen zu Schmerzen bei betroffenen Frauen. In isolierten, invasiven Epithelzellen (EEC145T) einer Endometriose-Läsion konnte die Expression von Shrew-1 gezeigt werden. Auch in anderen zellulären Zusammenhängen fördert die Expression von Shrew-1 den invasiven Phänotyp. Shrew-1 ist ein Transmembranprotein, das in Epithelzellen mit den Adhärenzverbindungen assoziiert ist und Interaktionen mit β-Catenin und E-Cadherin eingeht. In MCF7-Zellen fördert die Expression von Shrew-1 die EGF-induzierte Internalisierung von E-Cadherin, welche zur Verminderung der Zell-Zell-Adhäsion führt. In 12Z- und HT1080-Zellen konnte eine Interaktion mit CD147 gezeigt werden. CD147 fördert die Aktivität von MMPs und in Shrew-1-überexprimierenden HT1080-Zellen konnte eine erhöhte Aktivität der MMP9 gezeigt werden. Shrew-1 wirkt somit auf die Invasivität von Zellen und ist gleichzeitig Teil der Adhärenzverbindung. Aus diesem Grund wird Shrew-1 eine modulatorische Rolle in diesem Kontext zugeschrieben.
In immunhistologischen Färbungen von Shrew-1 und E-Cadherin konnte in Adenomyose-Läsionen eine inverse Expression der beiden Proteine in einigen epithelialen Zellen gezeigt werden, die im Endometrium nicht detektiert werden konnten. In den epithelialen Endometriose-Zelllinien 12Z und 49Z, die kein E-Cadherin exprimieren und äquivalent zu der Zelllinie EEC145T sind, führte die Herunterregulation von Shrew-1 (Shrew-1 KD) zur Reexpression von E-Cadherin. E-Cadherin ist in den 12Z Shrew-1 KD-Zellen an der Plasmamembran lokalisiert und interagiert mit β-Catenin, wodurch seine Assoziation mit den Adhärenzverbindungen wahrscheinlich ist. Die Herunterregulation von Shrew-1 führt zu einer verminderten Motilität und Invasivität der 12Z-Zellen, wobei die reduzierte Invasivität nicht alleine auf die Reexpression von E-Cadherin zurückgeführt werden kann. Es ist zu vermuten, dass das verminderte invasive Verhalten mit der ausbleibenden Interaktion von Shrew-1 mit CD147 zusammenhängt, welches die Aktivität von MMPs fördert.
Da Shrew-1 eine direkte Interaktion mit β-Catenin eingehen kann, ist es möglich, dass die Herunterregulation von Shrew-1 zu Veränderungen in der Lokalisation von β-Catenin und weiteren Proteinen, die mit den Adhärenzverbindungen assoziiert sind (p120 Catenin und Aktin), führen. Dies konnte jedoch nicht beobachtet werden. Eine verstärkte Lokalisation von Vinculin an den Enden von Aktin-Stressfasern sowohl in Zellausstülpungen als auch an Zell-Zell-Kontakten konnte in 12Z-Zellen nach der Herunterregulation von Shrew-1 beobachtet werden. Dies könnte eine Folge der E-Cadherin-Reexpression oder entscheidend für die Lokalisation von E-Cadherin an der Membran sein.
Die Reexpression von E-Cadherin, die in den 12Z Shrew-1 KD-Zellen auf mRNA- und Protein-Ebene nachgewiesen werden kann, erfolgt in den 12Z-Zellen vermutlich hauptsächlich über Veränderungen von Histon-Acetylierungen, da die Behandlung mit dem HDAC-Inhibitor TSA die Expression von E-Cadherin in den 12Z-Zellen induziert. Eine verstärkte H3K9-Acetylierung am CDH1-Promotor konnte in ChIP-Analysen in den 12Z Shrew-1 KD-Zellen gezeigt werden. Die gesteigerte Acetylierung resultiert vermutlich aus der verminderten Assoziation von HDAC1 und HDAC2 mit dem CDH1-Promotor in diesen Zellen. Eine Beteiligung der Repressoren Snail, Slug, Twist und ZEB1 an der Reexpression von E-Cadherin in den 12Z Shrew-1 KD-Zellen konnte nicht gezeigt werden. Ebenso scheinen Veränderungen am Methylierungsstatus des CDH1-Promotors nach der Herunterregulation von Shrew-1 nicht zu erfolgen.
TSA induziert auch in weiteren epithelialen Endometriose-Zelllinien (10Z und 49Z) die Expression von E-Cadherin. In stromalen Zellen führt hingegen weder TSA noch die Herunterregulation von Shrew-1 zur Expression von E-Cadherin (17B, 18B und 22B). Dies weist darauf hin, dass die Herunterregulation von Shrew-1 über die Veränderungen von Histon-Acetylierungen wirkt und dass dieser Mechanismus in epithelialen Endometriose-Zellen entscheidend ist. In den stromalen Zellen muss die Expression von E-Cadherin über einen anderen und/oder weitere Mechanismen blockiert sein.
Auch der Wnt-Signalweg scheint an der Reexpression von E-Cadherin in 12Z-Zellen beteiligt zu sein. Die Inhibierung der GSK3β (LiCl und SB216763) führt zur Expression von geringen Mengen an E-Cadherin. In 12Z Shrew-1 KD-Zellen führt die Stabilisierung von Axin (XAV939) zur verminderten Expression von E-Cadherin. Dies lässt darauf schließen, dass Shrew-1 auch einen Einfluss auf den Wnt-Signalweg hat, was vor allem durch dessen Interaktion mit β-Catenin wahrscheinlich ist.
Terrestrische Säugetiere werden von unterschiedlichen Parasiten als Wirte genutzt. Dabei kann ihre Parasitenfauna je nach Art, Lebensweise, Verbreitung, Gesundheitszustand und Reproduktionsstatus des Wirts abweichen. Ein weiterer bestimmender Faktor, ist der Einfluss des Menschen in Form von Regulierungsmaßnahmen und Schaffung urbaner Lebensräume. Domestizierte Haustiere bzw. Nutztiere weisen daher in der Regel andere Parasiten auf als ihre wildlebenden Artgenossen. Gleichzeitig können sich sowohl Wildtiere als auch domestizierte Tiere und Menschen gegenseitig Parasitenarten teilen und wechselseitig aufeinander übertragen. Daraus resultierende Krankheiten werden als Zoonosen bezeichnet.
Insbesondere Fledermäuse (Unterordnung Microchiroptera) zeigen weltweit eine enorme Parasitendiversität, die noch weitgehend unerforscht ist. Ebenfalls Forschungsbedarf besteht für die Sandfloh-Gattung Tunga in Süd- und Mittelamerika in Hinblick auf ihr Wirtsspektrum, welches auch Menschen einschließt. Die Art Tunga penetrans und zahlreiche weitere Parasitenarten, parasitieren gleichzeitig auch bei Hunden. Daher stellen diese Wirte eine direkte Gesundheitsgefahr für Menschen in ihrer unmittelbaren Umgebung dar.
Die vorliegende Dissertation ist in kumulativer Form zusammengefasst und beinhaltet drei Einzelpublikationen sowie einen Reviewartikel.
Ziel war es, die Parasitendiversität von Hunden aus urbanen tropischen Gebieten und die Parasitendiversität des Großen Ameisenbären (Myrmecophaga tridactyla) mit Hilfe morphologischer und molekularbiologischer Methoden zu analysieren. Die jeweiligen Parasitenfaunen wurden in Hinblick auf die soziale bzw. solitäre Lebensweise der beiden Wirtsarten verglichen und ihr zoonotisches Potenzial bewertet.
Ein weiteres Ziel war die Zusammenfassung der Ektoparasitennachweise süd- und mittelamerikanischer Microchiroptera und für die europäischen Arten der Fledermaus-Gattung Myotis (hier Endo- und Ektoparasiten) auf Basis der verfügbaren Literatur. Des Weiteren sollten eigene Parasitennachweise aus Bolivien bzw. Deutschland erfolgen. Für die Nachweise aus Deutschland wurden M. myotis untersucht, deren Artzugehörigkeit vorher bestimmt wurde. Zusätzlich wurden diese Individuen auf humanpathogene Lyssaviren untersucht.
Die Nachweise erfolgten über molekularbiologische und morphologische Methoden.
Vascular tumors associated with chronic B. henselae infections are unique examples of infection-associated pathological angiogenesis. The chaotic vascular architecture and prominent myeloid infiltrate of B. henselae induced vascular lesions show many similarities with malignant tumors.
In human cancers infiltrating myeloid cells play a decisive role in tumor progression and vascularization. In particular, tumor associated macrophages (TAMs) transform the tumor microenvironment, drive tumor invasion and vascularization through secretion of pro-angiogenic and immune modulatory cytokines and participation in matrix remodeling processes.
Myeloid angiogenic cells (MACs) are a subset of circulating myeloid progenitors with important roles in regenerative and pathological angiogenesis and a critical involvement in tumor vascularization. The phenotypic plasticity and importance of MACs in pathological angiogenic processes, position these cells as key potential players in B. henselae associated vascular tumor formation.
To investigate the possible role of MACs in B henselae induced pathological angiogenesis, the objective of this study was to examine the interaction of B. henselae with MACs and determine how this may affect their angiogenic capacity.
Building on previous work by Mӓndle (2005) this study has demonstrated that MACs are susceptible to infection with B. henselae and reside in intracellular vacuoles. As in endothelial cells, infection of MACs with B. henselae was associated with inhibition of apoptosis and activation of endogenous angiogenic programs including activation of the angiogenic transcription factor HIF-1.
In addition to angiogenic re-programming on a molecular level B. henselae infection increases MAC functional angiogenic capacity. B. henselae infected MACs were found to integrate into growing endothelium and increase the rate of angiogenic sprouting in a paracrine manner.
When cultured in a Matrigel capillary formation assay, infected MACs were also found to form networks of capillary-like structures that were stable over long periods of time. The B. henselae pathogenicity factor BadA was essential for the induction of this vascular mimicry phenotype as well as the activation of HIF-1 in infected MACs indicating that this factor may play an important role in MAC angiogenic re-programming.
Examination of infected MACs via FACS analysis, cytospin immunohistochemistry and qRT-PCR revealed that endothelial differentiation does not play a role in the B. henselae induced pro-angiogenic phenotype. Instead, MACs were shown to be myeloid in phenotype displaying typical macrophage markers which were upregulated upon B. henselae infection and maintained over long-term culture.
The increased angiogenic activity of B. henselae infected MACs was found to be associated with a broad phenotypic reprogramming in infected cells. In particular, gene expression programs related to angiogenesis, structural organization, apoptosis, sterol metabolism and immune regulation, were upregulated. Further examination of microarray gene expression profiles revealed that B. henselae infected MACs display a predominantly M2 anti-inflammatory macrophage activation status.
Finally, examination of the paracrine microenvironment created by B. henselae infected MACs revealed a diverse cytokine secretion profile dominated by inflammatory-angiogenic cytokines and matrix remodeling elements and lacking expression of some of the most important cytokines involved in the expansion of the inflammatory response. This B. henselae induced activation status was demonstrated to be distinct from the general inflammatory response induced by E. coli LPS treatment.
Comparison of B. henselae infected MACs to TAMs revealed many parallels in functional and phenotypic characteristics. Both TAMs and B. henselae infected MACs demonstrate increased angiogenic capacity, invasive, and immune modulatory phenotypes and the ability to participate in the formation of vascular mimicry phenotypes under angiogenic pressure. Furthermore, the pro-angiogenic paracrine microenvironment created by B. henselae infected MACs shows many similarities to the TAM-created tumor-microenvironment.
In conclusion, these investigations have demonstrated that the infection of MACs with B. henselae results in the phenotypic re-programming towards TAM-like cells with increased pro-angiogenic, invasive and immune-modulatory qualities. The results of this study elucidate new aspects of B. henselae pathogenicity in myeloid cells and highlight the role of these cells as paracrine mediators of B. henselae induced vascular tumor formation. In addition, these findings demonstrate that manipulation of myeloid cells by pathogenic bacteria can contribute to microenvironmental regulation of pathological tissue growth and suggest parallels underlying bacterial infections and cancer.
To overcome poor treatment response of pediatric high-risk acute lymphoblastic leukemia (ALL), novel treatment strategies are required to reactivate programmed cell death in this malignancy. Therefore, we take advantage of using small-molecule antagonists of Inhibitor of apoptosis (IAP) proteins, so called Smac mimetics such as BV6, which are described to overcome apoptosis resistance and thereby sensitize tumor cells for several apoptotic stimuli. To address the question whether redox alterations can sensitize leukemic cells for Smac mimetic-mediated cell death, we interfered with the cellular redox status in different ALL cell lines. Here, we show for the first time that redox alterations, mediated by the glutathione depleting agent Buthioninesulfoximine (BSO), prime ALL cells for BV6-induced apoptosis. Besides ALL cell lines, BV6/BSO cotreatment similarly synergizes in cell death induction in patient-derived primary leukemic samples. In contrast, the combination treatment does not exert any cytotoxicity against peripheral blood lymphocytes (PBLs) or mesenchymal stroma cells (MSCs) from healthy donors, suggesting some tumor selectivity of this treatment. We also identify the underlying molecular mechanism of the novel synergistic drug interaction of BSO and BV6. We demonstrate that both agents act in concert to increase reactive oxygen species (ROS) production, lipid peroxidation and finally apoptotic cell death. Enhanced ROS levels in the combination treatment account for cell death induction, since several ROS scavengers, like NAC, MnTBAP and Trolox attenuate BSO/BV6-induced apoptosis. BSO/BV6-induced ROS can be mainly classified as lipid peroxides, since the vitamin E derivate α-Tocopherol as well as Glutathione peroxidase 4 (GPX4), which both specifically reduce lipid-membrane peroxides, prevent lipid peroxidation, caspase activation and cell death induction. Vice versa, GPX4 knockdown and pharmacological inhibition of GPX4 by RSL3 or Erastin enhance BV6-induced cell death. Importantly, cell death induction critically depends on the formation of a complex consisting of RIP1/FADD/Caspase-8, since all complex components are required for ROS production, lipid peroxidation and cell death induction. Taken together, we demonstrate that BSO and BV6 cooperate to induce ROS production and lipid peroxidation which are eventually required for caspase activation and cell death execution. Collectively, findings of this study indicate that BV6-induced apoptosis is mediated via redox alterations offering promising new treatment strategy to overcome apoptosis resistance in ALL.
Hepatocellular carcinoma (HCC) is the fifth most common malignant tumor and third leading cause of cancer-related death worldwide. Most cases arise as a consequence of underlying liver disease, e.g. developed from chronic hepatitis B or C infectionsalcohol abuse or obesity, and are most often associated with liver cirrhosis. Hypoxiand the hypoxia inducible factors (HIF)-1α and -2α promote tumor progression of HCC, not only affecting tumor cell proliferation and invasion, but also angiogenesis and lymphangiogenesis and thus, increasing the risk of metastasis.
HCC is characterized as one of the most vascularized solid tumors. While HIF-1α and HIF-2α are frequently up-regulated in HCC only HIF-2α is correlated with high patientlethality. HIF-dependent regulation of HCC angiogenesis is controversially discussed.VEGFA, for example, as the most prominent factor inducing tumor angiogenesis represents not only a HIF-1 target, but also a HIF-2 target gene in HCC. This questions whether both isoforms have overlapping functions in regulating the angiogenic switch in HCC.
Besides angiogenesis also tumor-associated lymphangiogenesis significantly influences patient survival in HCC. Lymphatic spread is an important clinical determinant for the prognosis of HCC, but little is known how lymphangiogenesis is controlled in this context. To date, mainly HIF-1α was positively correlated with olymphatic invasion and metastasis in HCC, while a defined role of HIF-2α is missing. Thus, although HIF-1α and HIF-2α are structurally alike and regulate overlapping but not identical sets of target genes, they promote highly divergent outcomes in cancer progression and may even have counteracting roles. The aim of my work was to characterize the specific role of HIF-1α and HIF-2α in the angiogenic switch and lymphangiogenesis induction during HCC development.
Therefore, I created a stable knockdown of HIF-1α and HIF-2α in HepG2 cells and generated cocultures of HepG2 spheroids and embryonic bodies derived from embryonic mouse stem cells as an in vitro tumor model mimicking the cancer microenvironment to analyze which HIF isoform has key regulatory functions in HCC (lymph)angiogenesis. In cocultures with a HIF-2α knockdown angiogenesis was attenuated but lymphangiogenesis increased, while the knockdown of HIF-1α was without effect. Microarray analysis identified plasminogen activator inhibitor 1 (PAI-1)and insulin-like growth factor binding protein 1 (IGFBP1) as HIF-2 target genes.However, prominent angiogenic and lymphangiogenic factors such as VEGFs, PDGFB, ANG and their receptors were not regulated in a HIF-dependent manner. As PAI-1 was linked to angiogenesis in literature and IGF-signaling, which is negatively regulated by IGFBP-1, was correlated with lymphangiogenesis, I decided to investigate their HIF-2α-dependent influence on HCC (lymph)angiogenesis. The knockdown of PAI-1 in HepG2 cells also lowered angiogenesis in PAI-1k/d cocultures similar to the HIF-2α k/d phenotype. PAI-1 as the potent inhibitor of tPA and uPA, both inducing the conversion of plasminogen to plasmin, also inhibits plasmin directly. Therefore, I assumed an increase of plasmin in HIF-2α k/d and PAI-1 k/d cocultures as a result of the reduced PAI-1 levels. Blocking plasmin with aprotinin in HIF-2α k/d cocultures restored angioge nesis, suggesting that HIF-2α increases PAI-1 to lower concentrations of active plasmin, thereby supporting angiogenesis. In further experiments I could exclude PAI-1 to reduce angiogenesis by inducing plasmin-mediated apoptosis of differentiating stem cells in PAI-1 k/d and HIF-2α k/d cocultures, but demonstrated an increase of VEGFA165 degradation in these cocultures, suggesting plasmin-catalyzed proteolysis of VEGF as an additional layer of regulation required to explain the angiogenic phenotype. Besides the pivotal role of PAI-1 in angiogenesis I also investigated its potentialinfluence in lymphangiogenesis. Indeed, the knockdown of PAI-1 reduced lymphaticstructures and implied an important but opposing role in lymphangiogenesis comparedto induced lymphangiogenesis in HIF-2α k/d cocultures. However, blocking plasmin again with aprotinin in HIF-2α k/d cocultures restored lymphangiogenesis to the level of control virus, which indicates a divergent lymphangiogenic role of plasmin in PAI-1 k/d and HIF-2α k/d cocultures, possibly because of other essential pathways masking the lymphangiogenic effects of PAI-1 in HIF-2α k/d cocultures.
HIF-2α resulting in reduced IGFBP1 expression induced the differentiation of stem cells toward a lymphatic cell type and significantly enhanced the assembly of human dermal lymphatic endothelial cells into tubes. These data point the first time to an important impact of HIF-2 in the regulatin of lymphangiogenesis in vitro by inducing IGFBP1 and thus, scavenging IGF-1. Furthermore, matrigel plug assays to investigate the in vivorelevance of these observations confirmed HIF-2α as a crucial factor in the regulation of lymphangiogenesis in vivo
In conclusion, this work provides evidence that HIF-2α is a key regulator of angiogenesis and lymphangiogenesis in HCC by regulating PAI-1 and IGFBP1. HIF-2α positively influences the angiogenic switch via PAI-1 and negatively affects lymphangiogenesis via IGFBP1 expression. Targeting HIF-2α in HCC to reduce tumor angiogenesis should be approached carefully, as it might be overcome by induced lymphangiogenesis and metastasis.
Die Substitution von klassischen, mit der Nahrungsmittelproduktion in Konkurrenz stehenden, Substraten wie Glukose durch alternative Kohlenstoffquellen in der Biotechnologie ist sowohl aus ethischer, als auch aus ökonomischer Sicht erstrebenswert. Diese Arbeit beschreibt die Synthese von Bulkchemikalien in Form zweier Dicarboxylsäuren und einer Feinchemikalie in Form eines Sesquiterpens aus dem alternativen Substrat Methanol mit Hilfe genetisch veränderter Stämme des methylotrophen α-Proteobakteriums Methylobacterium extorquens.
Mesacon- und (2S)-Methylsuccinsäure sind Dicarboxylsäurederivate der CoA-Ester Mesaconyl- und (2S)-Methylsuccinyl-CoA, die als Intermediate im Ethylmalonyl-CoA- Weg (EMCP) vorkommen. M. extorquens nutzt den EMCP für die Regeneration von Glyoxylat, das für das Wachstum auf C1-Substraten wie Methanol obligatorisch ist. In dieser Arbeit konnte erstmals Mesacon- und (2S)-Methylsuccinsäure de novo durch die Expression einer für die Vorstufen Mesaconyl- und (2S)-Methylsuccinyl-CoA aktiven Thioesterase produziert werden. Ein kobaltlimitiertes Wachstum von M. extorquens führte aufgrund mangelnder Cofaktorversorgung zweier Vitamin-B12-abhäniger Mutasen im EMCP zu einer Akkumulation der beiden CoA-Ester-Vorstufen, womit eine Produktion von 0.65 g/l Mesacon- und (2S)-Methylsuccinsäure erreicht wurde. Weitergehende Untersuchungen belegten außerdem einen positiven Effekt eines ausgeschalteten PHB-Zyklusses auf die Produktion der beiden EMCP- Dicarboxylsäurederivate.
Diese Arbeit beinhaltet zusätzlich grundlagenwissenschaftliche Untersuchungen zur Substitution der EMCP-katalysierten Glyoxylatregeneration durch einen heterologen Glyoxylatzyklus in EMCP-negativen M. extorquens-Stämmen. Dabei konnte erstmals ein methanolverwertendes, methylotrophes Bakterium identifiziert werden, das einen Serin-Zyklus in Kombination mit dem Glyoxylat-Zyklus zur Kohlenstoffassimilation verwendet, ohne dabei zusätzliche Stoffwechselwege zur CO2-Fixierung wie den EMCP, RuMP oder CBB-Zyklus zu verwenden.
Die Präsenz einer nativen C30-Carotinoidbiosynthese, ausgehend von der Vorstufe Farnesylpyrophosphat (FPP), empfiehlt M. extorquens als Produktionsorganismus für (Sesqui-)Terpene. In dieser Arbeit wurde mit Hilfe einer induzierbar gesteuerten Expression einer Terpensynthase in Form einer α-Humulen-Synthase, einer FPP-Synthase und eines prokaryontischen Mevalonatweges, erstmals die de novo Synthese eines Terpens aus Methanol am Beispiel des α-Humulens etabliert. Durch optimierte Expressionen der Terpensynthase, FPPS und einzelner MVA-Gene mit Hilfe angepasster Translationsinitiationsraten der jeweiligen ribosomalen Bindestellen und der Verwendung eines in der nativen Carotinoidbiosynthese inhibierten M. extorquens-Stammes wurden finale Produkttiter von bis zu 1.65 g/l α-Humulen in Fed-Batch-Fermentationen erreicht.
Diese kumulative Dissertation beinhaltet außerdem einen Reviewartikel, in dem der verwendete Mikroorganismus M. extorquens in mikrobiologischer, genetischer, biochemischer und auch biotechnologischer Hinsicht ausführlich beschrieben wird. Zudem gibt ein Buchkapitel eine Übersicht über die Verwendung von Methanol in der Biotechnologie.
The RAF family of kinases constitutes the members A, B and CRAF. They mediate RAS signaling by linking it to the MEK/ERK transduction module, which regulates cellular processes such as cell proliferation, migration, survival and cell death. As the RAS/RAF/MEK/ERK (MAPK) pathway is found to be activated in human cancers, the RAF kinases have been exploited as valuable therapeutic targets and RAF inhibitors show promising results in the clinic, esp. with tumors harboring an activating BRAFV600E mutation. However, RAF inhibitors paradoxically accelerate metastasis in RAS mutant and BRAF wildtype tumors. They also become ineffective over time in BRAFV600E tumors because of reactivation of downstream mitogen-activated protein kinase (MAPK) signaling by promoting RAF dimerization. Aims of the present work were 1) to investigate the role of ARAF kinase in the paradoxical activation of the enzymatic cascade by RAF inhibitors downstream of mutated RAS and 2) to study the consequences of the loss of ARAF function on signal transduction in vitro and in vivo (nude mice). We have engineered several cell lines that would allow the study of basal and RAF inhibitor induced effects on MAPK activation, tumor cell migration and invasion.
In summary, we were able to show that the RAF isoform ARAF has an obligatory role in promoting MAPK activity and tumor cell invasion in a cell type dependent manner. In these cell types, ARAF depletion prevented the activation of MAPK kinase 1 (MEK1) and extracellular signal-regulated kinase 1 and 2 (ERK1/2) and led to a significant decrease of protrusions growing out of tumor cell spheroids in a three-dimensional (3D) culture that were otherwise induced by BRAFV600E-specific or BRAF/CRAF inhibitors (GDC-0879 and sorafenib, respectively). RAF inhibitors stimulated homodimerization of ARAF and heteromerization of BRAF with CRAF and the scaffolding protein KSR1. However, induced oligomerization was not sufficient to activate MAPK signaling if ARAF was depleted. By employing full length recombinant kinases, we were able to show for the first time that the three RAF isoforms competed for the binding to MEK1. In cell culture models, the overexpression of dimer-deficient ARAF mutants impaired the interaction between ARAF and endogenous MEK1 and thus prevented the subsequent phosphorylation of MEK1 and ERK1/2. Our findings reveal a new role for ARAF in directly activating the MAPK cascade through homodimerization and thereby promoting tumor cell invasion, suggesting the conserved RAF-dimer interface as a target for RAS- and RAF mediated cancer therapy.
Collectively, we provide evidence for the dual role ARAF plays in controlling MAPK signaling and cancer as loss of ARAF promoted strong lung metastasis formation in nude mice. Preliminary data describing the underlying mechanisms behind ARAF-regulated metastases have been presented and discussed.
The brain vascular system is composed of specialized endothelial cells, which regulate the movement of ions, molecules and cells from the blood lumen to the central nervous system (CNS). Endothelial cells in the brain form the blood-brain barrier (BBB) that is essential to maintain the brain homeostasis and protect the CNS from pathogens and toxins for a proper neurological function. Endothelium together with other cellular components such as pericytes, astrocytes and the basement membrane, forms the neurovascular unit (NVU), the structural unit of the BBB. Breakdown of the BBB occurs in various neurological disorders, leading to edema and neuronal damage. Therapeutic strategies focusing on factors that regulate the permeability of the BBB may help to improve neurological disorders and facilitate drug delivery to the brain.
Angiopoietins (Ang) are potential candidates for therapeutic targeting the BBB due to their role in regulating the vascular permeability in periphery. They are key growth factors that control angiogenesis and vessel maturation. Ang-1 and Ang-2 possess similar binding affinities to the Tie2 receptor tyrosine kinase, which is almost exclusively expressed on endothelial cells. Ang-1 is expressed in smooth muscle cells and pericytes, and binds in a paracrine manner to Tie2. This results in phosphorylation of the receptor and induction of downstream signaling pathways leading to vessel maturation via pericyte recruitment and blood vessel stabilization. Ang-2, on the other hand, is stored in Weibel Palade bodies in endothelial cells and is released upon inflammatory or angiogenic stimuli. Therefore, in mature, stabilized blood vessels, Ang-2 expression is low. Increased level of Ang-2 is only observed during development or in pathology such as ischemia, cancer and inflammation. When Ang-2 is released, it acts in an autocrine manner and interferes with Tie2 phosphorylation in a context-dependent way. Antagonizing the receptor results in de-stabilization of the vessels, often accompanied by reduced numbers of pericytes leading to myeloid cell infiltration. Cyanobacteria belong to the most widely distributed microorganisms in the biosphere and contribute significantly to global primary production. Their metabolism is based on oxygenic photosynthesis and some cyanobacteria can fix elemental nitrogen. Obligate photosynthetic diazotrophs have a particularly high iron demand in comparison to heterotrophic bacteria. Nevertheless the understanding of iron acquisition in cyanobacteria is just beginning to emerge. Iron acquisition in bacteria comprises highly specific transport of siderophore-iron complexes over the outer membrane by TonB-dependent transporter (TBDT). The transport itself is active and energized by a multi-complex localized to the inner membrane termed the TonB-system (TonB-ExbB-ExbD). The siderophore-iron complexes are further transported into the cytosol by a binding protein dependent ABC-transporter. Cyanobacterial iron acquisition response has most extensively been studied in unicellular, non-siderophore synthesizing cyanobacteria in the genus Synechococcus and Synechocystis. Anabaena sp. PCC 7120, however, is a different model organism as it is a freshwater living, siderophore synthesizing and, truly multicellular microorganism. It can be assumed that siderophore synthesis and siderophore-dependent iron uptake are tightly coordinated processes, therefore Anabaena represents a different model organism as compared to non-siderophore producing cyanobacteria. Moreover the surprisingly abundant protein family of 22 putative TBDTs in Anabaena indicates a high complexity of TonB-dependent uptake systems. Sequence similarity analysis revealed 4 putative tonB encoding genes (alr0248, all3585, all5036, alr5329), 2 putative exbB-exbD encoding gene cluster (alr0643-alr0644, all5047-all5046), one single standing putative exbB encoding gene (alr4587) and several hypothetical binding-protein-dependent ATP binding cassette (ABC)-type transporter encoding genes (fhu-, fec- and fut-type transporter).
In this study the respond of the predeicted systems to iron-limiting conditions was analysed by qRT-PCR. The expression analysis revealed on the one hand an enhanced transcription of all5036 (tonB3), all5047-all5046 (exbB3-exbD3) and the fhu-like encoding genes (all0387-all0389) under iron-limitation and at the same time down-regulation of expression under enhanced iron concentrations. Summerizing the transcription profile of the tonB3- and the fhu-system showed an expression regulated by iron-availability. To further characterize the role of TonB3-, ExbB3- and the Fhu-system, mutants thereof were generated. None of the generated mutants, except for the exbB3 mutant, could be fully segregated, suggesting an essential character of the genes. Characterization of the mutants revealed enhanced expression of iron-starvatrion indicator genes (isiA, fhuA) and altered growth of the tonB3 mutant under iron-limiting conditions. The iron starvation phenotype was further strengthened by enhanced siderophore secretion in the tonB3, exbB3 and fhuC mutants. Taken as a whole the results strongly indicate involvement of the tonB3- and the fhu-system in siderophore-dependnet iron uptake in Anabaena.
Investigation of the tonB2 (all3585) mutant under iron and citric acid limitation resultated in altered growth of the mutant. However, growth could be restored by addition of iron chlorid. Therefore a connection of the TonB2 protein to iron uptake is implied and further supported by ressitance to toxic iron concentrations. Lastly, mutation of tonB1 (alr0248) reuslted in insensibility to toxic manganese and copper concentrations and macrolid antibiotics. The altered permeability of the outer membrane may be a result of decreased expression of seven putative porin encoding genes in the mutant. A possible role in transcriptional regulation of porin expression is discussed.
In der vorliegenden Doktorarbeit zur Untersuchung der Rolle der Superoxid-Dismutasen in P. anserina lieferten die durchgeführten Analysen folgende Ergebnisse:
1)Sowohl in P. anserina als auch in S. cerevisiae wurde eine gemeinsame Regulation von SODs nachgewiesen: Stämme, die die mitochondriale MnSod (PaSod3 bzw. ScSod2) überexprimieren zeigen eine erhöhte Cu/ZnSOD-Aktivität (PaSOD1 bzw. ScSOD1).
2)Es konnte keine SOD-Aktivität für die putativen SODs Pa_1_10620, Pa_1_10630 und Pa_1_6300 detektiert werden. Für Pa_1_10620, dessen Überexpression unter Standardbedingungen zu einer Lebensverlängerung führt, wird eine Funktion als mitochondriales ribosomales Protein angenommen.
3)Der ∆PaSod3-Stamm weist keinen Unterschied im Phänotyp, der Wuchsrate und der Lebensspanne unter Standardbedingungen zum Wildtyp auf. Paraquat-Stress führt allerdings zu einer Kurzlebigkeit des ∆PaSod3-Stammes, wohingegen diese Mutante eine höhere Resistenz gegenüber Wasserstoffperoxid aufweist als der Wildtyp.
4)Transkriptomanalysen des Wildtyps und der ∆PaSod3-Mutante lassen vermuten, dass eine Hochregulation von Detoxifizierungs- und Energie-abhängigen Prozessen die durch den Verlust der mitochondrialen PaSOD3 vermuteten negativen Auswirkungen kompensieren.
5)PaSod3_OEx-Stämme weisen unter Standardbedingungen aufgrund der erhöhten intrazellulären Wasserstoffperoxid-Menge, bedingt durch die vermehrte Umsetzung von Superoxid, diverse negative Auswirkungen auf: Eine reduzierte Wuchsrate, verkürzte Lebensspanne, geringere Fertilität, stärkere Pigmentierung, vermehrt fragmentierte Mitochondrien, mehr unprozessierte mitochondriale Proteine und weniger Komplex IV der Atmungskette als der Wildtyp. Zusätzlich wird vermehrt über die alternative Oxidase geatmet, um die ROS-Generierung zu reduzieren.
6)Oxidativer Stress in Form von Paraquat führt in PaSod3_OEx-Stämmen zu einer weiteren Verkürzung der medianen Lebensspanne, während die maximale Lebensspanne von PaSod3_OEx3-Stämmen im Vergleich zum Wildtyp sogar verlängert ist. Wasserstoff-peroxid resultiert in stark verringerten medianen und maximalen Lebensspannen beider PaSod3-überexprimierenden Stämme.
7)Die Anzucht auf Medium mit zusätzlichem Mangan (80 µM MnSO4) kann die beobachteten Defekte der PaSod3_OEx-Stämme fast vollständig auf Wildtyp-Niveau revertieren: Die Wuchsrate, die Lebensspanne, der Phänotyp, die Mitochondrien-morphologie, die Prozessierung mitochondrialer Proteine und die Atmung entsprechen dem Wildtyp. Lediglich die Fertilität erreicht nicht das Wildtyp-Niveau. Diese positiven Effekte von Mangan werden erzielt, da die erhöhte Wasserstoffperoxid-Menge in PaSod3_OEx-Stämmen entsprechend ihrer Entstehung detoxifiziert wird, denn Mangan führt zu einer gesteigerten Transkription bzw. Aktivität von Katalasen und Peroxidasen sowie zu einer erhöhten Peroxiredoxin-Menge.
8)Die Anzucht des Wildtyps unter Wasserstoffperoxid-Stress resultiert in einer Lebens-spannenverkürzung. Diese kann durch Supplementation mit Mangan revertiert werden. Unter diesen Bedingungen weisen u. a. Peroxidasen eine erhöhte Aktivität auf.
Insgesamt ließen die gewonnenen Daten den Schluss zu, dass das Genom von P. anserina für drei aktive SODs kodiert. Ein Verlust der einzigen mitochondrial lokalisierten SOD kann durch die Induktion von Energie-abhängigen Prozessen sowie von Detoxifizierungsprozessen kompensiert werden. Ferner weisen die durchgeführten Studien darauf hin, dass PaSod3_OEx-Stämme als Modell für erhöhte intrazelluläre Wasserstoffperoxid-Mengen in P. anserina verwendet werden können. Darüber hinaus wurde ein Zusammenhang zwischen Mangan und dem Detoxifizierungs-netzwerk in P. anserina nachgewiesen. Dabei können zwei Mechanismen zur Reduktion der Wasserstoffperoxid-Mengen unterschieden werden: Bei Vorhandensein ausreichender Mengen Mangan kommt es zu einer stärkeren Detoxifizierung von Wasserstoffperoxid. Ist Mangan allerdings limitiert und die Detoxifizierung kann nicht gesteigert werden, wird eine Umstellung der Atmung eingeleitet, um die neu entstehende ROS-Menge zu minimieren.
Evaluierung der zellfreien Produktion sekundär aktiver Transporter für die Proteinkristallisation

The four subunit (SU) aa3 cytochrome c oxidase (CcO) from Paracoccus denitrificans is one of the terminal enzymes of the respiratory chain. It uses electrons from cytochrome c to reduce molecular oxygen to water. Its binuclear active center, residing in SU I, contains hemeÊa3 and CuB, the latter being liganded by three histidine residues. Apart from its oxygen reductase activity, the protein possesses a peroxidase and a catalase activity.
To compare variants and the wild type (WT) protein in a more stringent way, a recombinant (rec.) WT CcO was constructed, carrying the gene for SUÊI on a low copy number plasmid. This rec. WT showed, as expected, no difference in oxygen reductase activity compared to the American Type Culture Collection (ATCC) WT CcO but surprisingly its catalase activity was increased by a factor of 20. The potential overproduction of SUÊI due to plasmid coding and the resulting deficiency in metal inserting chaperones might impair the correct insertion of hemeÊa3 and CuB because of a deficiency in metal inserting chaperones. This in turn might lead to differences in side chain orientation and to changes in the water network. However, slight changes might cause an increased accessibility of the active center for hydrogen peroxide, resulting in an increased catalase activity. The availability of chaperones and therefore the proposed structural reasons for the difference was improved by cloning the genes for the two metal inserting chaperones CtaG and Surf1c on the same plasmid together with SUÊI. This new rec. WT CcO showed in fact a reduced catalase activity. Another WT with a deletion in the chromosomal second, non expressing gene of SU I was analysed to prove plasmid coding as the reason for the difference of the ATCC WT and the rec. WT. This strain showed an increased kcat of the catalase activity as well, additionally pointing to a regulatory effect of the non expressed gene for SU I in the chromosome. To fathom the structural difference of the increased catalase activity, differential scanning calorimetry was used, but no significant difference in thermal stability between the ATCC WT CcO and the rec. WT CcO was detected. However, upon aging, the thermal stability of the rec. WT CcO declined faster than that of the ATCC WT CcO pointing to a decreased structural stability of the rec. WT CcO.
To characterize the catalase reaction, several known inhibitors were used to probe the contribution of the different metal cofactors in the catalase reaction. In addition variants in aromatic amino acids near the active center were constructed to conclude on a possible reaction mechanism of the catalase activity of CcO. These variants in combination with the wild type forms were analysed for radical signals by EPR-spectroscopy. A radical relevant for the catalase reaction of CcO was found in the F-intermediate of all variants and all wild type forms. This narrow 12 G radical signal was assigned to a porphyrine radical probably involved in the catalase reaction of CcO. Moreover, gas chromatography-mass spectrometry measurements were used to analyse isotopically labelled oxygen produced in the catalase reaction.
As a result of these experiments, a reaction cycle of the catalase activity of CcO is postulated and the structural difference between the ATCC and rec. WT CcO is outlined. The catalase activity appears to be a true catalase activity and not a "pseudocatalase" activity.
Snake bite envenoming often results in disability or death of breadwinners of poor families in the rural tropics and the subtropics of Nepal. Identification of the medically relevant snake species, circumstances of venomous snake bites, prehospital care of their bites and human responses to snakes and snake bite is, therefore, crucial to enable victims or first aider to select the appropriate first aid measures, physicians to anticipate complications and to use appropriate treatment protocols as well as the local community to implement prevention strategies. Inadequate educational gaps exist in Nepal and hinder identification of snakes involved in bites. To fill this gap, I aim to provide an evidence-based list of medically relevant snake species. Snake specimens brought by patients bitten or their attendants from the tropical and subtropical regions in southeastern, southcentral, and southwestern Nepal to snake bite treatment centres over a period from 2010 through 2014, were taxonomically identified and medical records of envenoming were evaluated.
In Nepal, the epidemiology of snake bite is poorly known. Here I describe the ecological circumstances of proven krait (Bungarus spp.) and Russell´s Viper (Daboia russelii) bites to elucidate and examine, whether environmental circumstances or human behaviour contributed to envenoming. In a cross-sectional study, data about prehospital care, environmental circumstances of 46 krait and 10 Russell´s Viper bites were evaluated. Patients were interviewed using structured interview forms. Snake bite prone communities were surveyed to test people´s knowledge on snakes and their attitude towards venomous snakes in general.
Of 349 snakes involved in bites, 199 (57%) specimens were found to be medically relevant venomous snakes that included 11 species belonging to six genera and two families. Among them, Naja naja (n = 76, 22%), Bungarus caeruleus (n = 65, 19%) and Trimeresusurs albolabris (n = 10, 3%) were the most widely distributed snakes. Daboia russelii (n = 10, 3%) was found to be restricted to the southwestern part of Nepal. For B. walli, a previously poorly known species, 13 voucher specimens represent the first country records of this species as well as the first documented cases of involvement in snake bite envenoming by this species in Nepal.
Numerous snake bites (33%) occurred at night, during the rainy season, and are mainly due to Bungarus species, particularly B. caeruleus. Bites of cobras and Russell’s Vipers are a risk at daytime. Evaluation of data regarding the place where the bite happened, indicates that the snake bite risks appear to be as high in residential areas, in and around houses, as in rural areas. In cases of kraits (n = 46), 61% of the bites occurred while the victim was sleeping indoors, those of Russell´s Vipers mainly during agricultural activities in the fields. Analysis of socio-demographic data revealed that both krait and viper bites predominantly affected farmers or their family members. However, snake bites involved also people of higher socio-economic status, which suggests that it is not a health problem of poor people only living in the rural areas of Nepal.
A small number of snake bite victims (n = 7) sought help from traditional healers, but most patients went to hospitals for medical treatment using motorbikes (65%) or were transferred by ambulance cars (22%). As a first aid measure, most patients (78%) had used a tourniquet, which is of doubtful value and has often severe sequelae, instead of applying the WHO recommended pressure immobilisation bandage or local compression pad. The overall case fatality rate was calculated to be 10%, but up to 17% in cases of Bungarus spp. bites.
Rural community people were found to be extremely afraid of snakes, a major reason for indiscriminate killing of even harmless snakes, e.g., Lycodon aulicus, which were wrongly considered to be venomous. This is mainly due to the poor knowledge on snakes in general and on their role in providing ecological services, which may eventually lead to a decline in snake populations and even the extinction of rare species.
The results of the present study strongly emphasize that snake bite is an important public health issue in Nepal. There is an urgent need to improve the knowledge of people on snakes and to try changing their attitudes towards these reptiles, in addition to documenting the biodiversity and distribution of medically relevant snakes, the epidemiology and circumstances of their bites. Avoiding high-risk behaviour (e.g., killing of snakes), using screened doors and windows are some of the suggested measures preventing snake bite. Early and accurate identification of the snakes involved should help physicians to apply timely treatment, eventually referring the patient to the appropriate hospital. This also has important implications in developing public health and conservation strategies, to the benefit of the people of Nepal.
In conclusion our data show, that Flightless I function is essential for striated muscle development in zebrafish. Myofibrillar bundling and focal adhesion formation represent the basis for this development, and are ultimately a prerequisite for cardiac trabeculation. Future analysis of Actin polymerization in trabeculation will provide addition knowledge about the sensitivity of the developing and adult heart to a disequilibrium in F-actin versus G-actin availability.
In this study we found a novel ErbB2-dependent cardiomyocyte maturation process which affects both cardiac chambers. It will be of great interest to further study the nature of the Memo1-GFP cell-cell junctions and other junction proteins in order to unravel the significance of this maturation process for heart development.
Interestingly we found, that memo1bns4 homozygous mutant animals, which we generated with CRISPR/Cas9 technology, develop indistinguishable from siblings, suggesting that zygotic memo1 expression is dispensable for zebrafish development. Future studies will address the question if maternal zygotic memo1bns4 mutants will develop a heart or vascular phenotype as reported form Memo1 knockout mice or as observed in memo1 morphants in this study.
In cultured C2 mouse skeletal muscle cells the Golgi-apparatus relocalizes dependent on centrosomal proteins and independent of microtubules. We describe here that zebrafish cardiomyocytes have a similar Golgi-complex distribution suggesting a similar differentiation-dependent reorganization. This striated muscle specific, fragmented Golgi distribution might be an advantage for these cells in order to shuttle vesicles through the densely packed sarcomere structures. Future studies could address the timing of the Golgi-reorganization in cardiomyocytes during development and possibly use this Golgi-zebrafish line as a tool to study cardiomyocyte maturation in disease models and in heart regeneration.
Stechmücken (Dipteren: Culicidae) sind weltweit mit über 3500 Arten und mit Ausnahme der arktischen Regionen ubiquitär vertreten. Die medizinische Relevanz dieser Tiergruppe, begründet durch die hämatophage Lebensweise der Weibchen, erschloss sich bereits Ende des 19. Jh. und hat bis heute Bestand. Jedes Jahr sterben rund 600.000 Menschen an den Folgen der Malaria und fast 100 Mio. Menschen infizieren sich mit dem Denguefieber. Zwar beziehen sich diese Zahlen fast ausschließlich auf die Entwicklungsländer, aber im Zuge des Klimawandels und des immer stärkeren Welthandels kommt es auch in Europa und den USA immer wieder zu Ausbrüchen vorher nicht relevanter Krankheiten. So hat sich das West-Nil- Virus seit 1999 in Nordamerika rasant verbreitet. Im Jahr 2013 gab es dort rund 2500 Fälle, von denen 119 zum Tod führten. In Europa traten hingegen Krankheiten wie das Chikungunyafieber (Italien 2007) oder das Denguefieber (Frankreich 2010/2013) auf. Die Gründe für diese Ausbrüche sind vor allem in der Einschleppung neuer Vektorspezies und Krankheitserreger sowie in den veränderten Wirtspräferenzen einheimischer Stechmückenarten zu suchen. Das Wissen um das Vektorpotential der in Deutschland heimischen Stechmücken konnte vor allem durch die seit 2009 initiierten Monitoring-Programme stetig erweitert werden. Auch die Veränderung der heimischen Fauna durch invasive Arten wie Ochlerotatus japonicus japonicus oder Aedes albopictus wird intensiv erforscht. Dennoch ist hinsichtlich der Biologie, Ökologie sowie Genetik vieler Arten noch immer wenig bekannt.
Die vorliegende Dissertation, welche auf Basis von vier (ISI-) Einzelpublikationen kumulativ angefertigt wurde, beschäftigte sich mit der Analyse der genetischen Variabilität sowie der Zoogeographie der untersuchten Arten und der Etablierung einer schnellen und kostengünstigen Methode zur Artdiagnostik. Besonderes Augenmerk wurde bei den Analysen auf die beiden heimischen Arten Culex pipiens und Culex torrentium sowie die invasive Art Ochlerotatus japonicus japonicus gelegt. Ziel war es, die noch bestehenden Wissenslücken zu füllen, um zukünftige Monitoring-Programme besser koordinieren sowie Analysen zur Vektorkompetenz und Genetik dieser Arten gezielter durchführen zu können.
Es konnte gezeigt werden, dass Cx. pipiens und Cx. torrentium deutliche Unterschiede in ihren Populationsstrukturen aufwiesen welche auf verschiedene evolutive Prozesse hindeuten. Die geringere genetische Variabilität in Cx. pipiens lässt auf positive Selektion durch z.B. Insektizidresistenz im Zuge durchgeführter Bekämpfungsmaßnahmen oder die Infektion mit Wolbachien schließen. Die analysierte Populationsstruktur von Cx. torrentium spricht hingegen für eine geringe Ausbreitung, wodurch der genetische Austausch reduziert wurde und so die untersuchten Populationen genetisch stärker voneinander abwichen. Des Weiteren ließen die Analysen des Cytochrom c Oxidase Untereinheit 1-Fragmentes (cox1) Rückschlüsse auf die Zoogeographie dieser Arten in Deutschland zu - wobei beide Arten über das Untersuchungsgebiet verteilt waren, Cx. torrentium jedoch in den neuen Bundesländern weniger häufig nachgewiesen wurde als in den alten und eine geringere gefangene Individuenzahl aufwies. Basierend auf der ökologischen Nischenmodellierung konnten potentiell neue Verbreitungsgebiete für die Art Ochlerotatus japonicus japonicus identifiziert werden. Als klimatisch besonders günstig zeigten sich dabei Südhessen, das Saarland sowie nördliche Teile Nordrhein-Westfalens. Mit Hilfe der etablierten Methode der direct-PCR wird in Zukunft eine schnellere und kostengünstigere Identifizierung von Stechmücken erfolgen können, welche aufgrund bestimmungsrelevanter Merkmale nicht mehr morphologisch zu identifizieren sind.
Um das Wissen über die Stechmücken in Deutschland fortlaufend zu intensivieren, ist sowohl das Weiterführen der Monitoring-Programme als auch die molekularbiologische Aufarbeitung der Proben nötig. Durch die Anwendung neuer Techniken und weiterer molekularer Marker wird es möglich sein, weitere Krankheitserreger sowie genetische Besonderheiten der heimischen Stechmückenfauna nachzuweisen. Aber auch die Überwachung invasiver Stechmückenarten durch die Modellierung potentieller Verbreitungsgebiete und die Anwendung molekularbiologischer Analysemethoden zum Detektieren der Arten und möglicher Krankheitserreger wird ein wichtiger Bestandteil der weiteren Forschung sein.
