Lesion of the rat entorhinal cortex denervates the outer molecular layer of the fascia dentata followed by layer-specific axonal sprouting of uninjured fibers in the denervated zone. One of the candidate molecules regulating the laminar-specific sprouting response in the outer molecular layer is the transmembrane chondroitin sulfate proteoglycan NG2. NG2 is found in glial scars and has been suggested to impede axonal regeneration following injury of the spinal cord. The present study adressed the question whether NG2 could also regulate axonal growth in denervated areas of the brain. Therefore, (1) changes in NG2 mRNA and NG2 protein levels, (2) the cellular and the extracellular localisation of the molecule, (3) the identity of NG2 expressing cells, and (4) the generation of NG2-positive cells were studied in the rat fascia dentata before and following entorhinal deafferentation. Laser microdissection was employed to selectively harvest the denervated molecular layer and combined with quantitative reverse transcription-PCR to measure changes in NG2 mRNA amount (6h, 12h, 2d, 4d, 7d post lesion). The study revealed increases of NG2 mRNA at day 2 (2.5-fold) and day 4 (2-fold) post lesion. Immunocytochemistry was used to detect changes in NG2 protein distribution (1d, 4d, 7d, 10d, 14d, 30d, 6 months post lesion). NG2 staining was increased in the denervated outer molecular layer at 1 day post lesion, reached a maximum at 10 days post lesion, and returned to control levels within 6 month. Interestingly, the accumulation of NG2 protein was strongly restricted to the denervated outer molecular layer forming a border to the unaffected inner molecular layer. Using electron microscopy, NG2-immunoprecipitate was localized not only on glial surfaces and in the extracellular matrix but also in the vicinity of neuronal profiles indicating that NG2 is secreted following denervation. Double-labelings of NG2-immunopositive cells with markers for astrocytes, microglia/macrophages, and oligodendrocytes suggested that NG2-cells are a distinct glial subpopulation before and after entorhinal deafferentation. Bromodeoxyuridine-labeling revealed that some of the NG2-positive cells are postlesional generated. Taken together, the data revealed a layer-specific upregulation of NG2 in the denervated outer molecular layer of the fascia dentata that coincides with the sprouting response of uninjured fibers. This suggests that NG2 could regulate lesion-induced axonal growth in denervated areas of the brain.
Die Dissertation liefert einen Beitrag zur Identifizierung und Charakterisierung der an der Komplementresistenz von Borrelien beteiligten CRASP-Proteine aus Isolaten der Genospezies B. burgdorferi s.s. und B. afzelii. Im Rahmen der Arbeit gelang es mittels Identifizierung und immunologischer Charakterisierung die Zugehörigkeit der spezifisch Faktor H-bindenden BbCRASP-Proteine BbCRASP-3, BbCRASP-4 und BbCRASP-5 zur Erp-Proteinfamilie zu beweisen. Weiterhin konnten die Faktor H- und FHL-1-bindenden BbCRASP-Proteine BbCRASP-1 und BbCRASP-2 von B. burgdorferi s.s. identifiziert werden. Mit dem BbCRASP-2-Protein wurde ein bis dahin unbekanntes Faktor H- und FHL-1-bindendes CRASP-Protein aus den äußeren Membranen des B. burgdorferi s.s.-Isolates B31 isoliert und charakterisiert. BbCRASP-2 stellt innerhalb der CRASP-Proteinfamilie ein neues eigenständiges Lipoprotein dar und unterscheidet sich deutlich von den Sequenzen der anderen CRASP-Proteine. Es ist weder ein Mitglied der gbb54- oder der Erp-Proteinfamilie, noch gehört es zu einer anderen bekannten Proteinfamilie von B. burgdorferi s.s. In Ligandenaffinitätsblot-Analysen konnte mit Hilfe von rekombinantem FHL-1 sowie Deletionsmutanten von Faktor H und FHL-1 gezeigt werden, dass die Bindung von Faktor H und FHL-1 an das BbCRASP-2-Protein ausschließlich über die SCR 7-Domäne vermittelt wird. Die Analysen C terminaler Deletionsmutanten von BbCRASP-2 unterstrichen die Bedeutung der letzten 16 Aminosäuren des BbCRASP-2-Proteins für die Interaktion mit Faktor H und FHL-1.
An die Signalübertragung im ZNS werden in bestimmten Entwicklungsstadien sehr unterschiedliche Anforderungen gestellt. Im adulten Gehirn dient sie nicht nur der Nachrichtenübermittlung, sondern auch der aktivitätsbasierten Umgestaltung neuronaler Verbindungen bei Lernprozessen und der regenerativen Umgestaltung nach Verletzungen. Im sich entwickelnden Gehirn werden schon frühzeitig Nervenzellen elektrisch erregbar und spontan aktiv. Diese elektrische Aktivität und die dadurch verursachte Transmitterausschüttung spielen bei der selektiven Stabilisierung von Synapsen und damit bei der Ausgestaltung des neuronalen Netzwerks eine große Rolle. Im ZNS von Vertebraten beruht die Wirkung des Neurotransmitters und Neuromodulators Acetylcholin auf den nikotinischen und muskarinischen Acetylcholinrezeptoren. Muskarinische Acetylcholinrezeptoren (mAChRen) koppeln, abhängig von den fünf identifizierten Rezeptorsubtypen (M1- M5), an unterschiedliche intrazelluläre Signalketten an. Dabei interagieren für gewöhnlich die M1, M3 und M5 Rezeptoren mit einem G-Protein des Typs Gq/11, während M2 und M4 ein G-Protein des Typs Gi/o aktivieren. Der Colliculus inferior (IC) ist eine wichtige Verschaltungsstation im auditorischen Mittelhirn von Säugetieren. Inhibitorische und exzitatorische Eingänge werden dort während der Entwicklung mit großer Präzision angelegt und konvergieren auf einzelne ICNeurone. Die physiologische Bedeutung von mAChRen im IC ist weitgehend unerforscht und die Subtypen die im juvenilen IC eine Rolle spielen wurden noch nicht charakterisiert. Es war das Ziel der vorliegenden Arbeit mittels elektrophysiologischer Untersuchungen im IC der juvenilen Ratte (P5-P12) folgende Fragen zu klären: i) Gibt es im IC der jungen Ratte eine Modulation der GABAergen Transmission durch muskarinische Acetylcholinrezeptoren? ii) Welcher muskarinische Rezeptorsubtyp spielt dabei eine Rolle? iii) Welcher intrazelluläre Signalmechanismus ist der Aktivierung des muskarinischen Acetylcholinrezeptors nachgeschaltet? Unter Wirkung von Muskarin kam es bei 41,2% der untersuchten Neurone des Colliculus inferior zu einer Erhöhung der Frequenz der spontanen IPSCs. Die sIPSCs wurden durch Bicucullin blockiert, somit handelt es sich um GABAerge IPSCs. Die Wirkung von Muskarin nahm zu, wenn die Tiere älter als 9 Tage waren. Es wurde gezeigt, dass nAChRen keine Rolle spielen bei der Erhöhung der sIPSC-Frequenz, während eine selektive Blockade der M3-(M5-) mAChRen durch 4-DAMP die Muskarinwirkung blockierte. In der Regel sind die M3-Rezeptoren über folgende Signalkaskade aktiv: Phospholipase C, Kalzium-Calmodulin, NO-Synthase, Guanylatzyklase, die NO-Konzentration und cGMP. Alle Glieder dieser Kaskade wurden untersucht, sie hatten aber keinen Einfluss auf die Muskarinwirkung. Im Gegensatz dazu führte die Erhöhung des intrazellulären cAMP-Spiegels zu einer vergleichbaren Frequenzsteigerung der sIPSCs wie sie unter der Wirkung von Muskarin gemessen wurde. Weiterhin ließ sich die Erhöhung der sIPSC-Frequenz durch Muskarin vollständig aufheben, wenn die intrazelluläre Adenylatzyklase blockiert wurde. Es ist bekannt, dass der M3-(M5-) mAChR über verschiedenste Signalwege den intrazellulären cAMP-Spiegel verringern oder erhöhen kann und dass die Spezifizierung des Rezeptors vom Grad der Rezeptorexpression, dem Zelltyp und der Kombination der Effektormoleküle abhängig ist. Die Abweichung der juvenilen Kaskade vom im erwachsenen Tier üblichen Signalweg hat dabei den Vorteil, dass eine Aufgabentrennung zwischen Netzwerkbildung und Synapsenstabilisierung einerseits und Signalweiterleitung andererseits erfolgt. Beim juvenilen Tier steht die Netzwerkbildung im Vordergrund, beim erwachsenen Tier wird die Signalweiterleitung moduliert. Der sekundäre Botenstoff NO, der weit diffundieren kann, spielt beim juvenilen Signalweg keine Rolle und dies ermöglicht eine klare Trennung der aktivierten von den umgebenden Synapsen. Dadurch wird eine starke Selektion innerhalb der vorhandenen Synapsen ermöglicht und eine aktivitätsbasierte Netzwerkbildung vereinfacht. Die Etablierung funktioneller GABAerger Synapsen ist ausschlaggebend für die Entwicklung des neuronalen Netzwerks. Innerhalb der ersten postnatalen Wochen führt die Aktivierung von GABAA-Rezeptoren zur Depolarisierung der IC-Neurone und zu einer Erhöhung der intrazellulären Ca2+-Konzentration. Acetylcholin potenziert also letztlich den GABA-aktivierten Einstrom von Ca2+ und führt damit zur Erhöhung der intrazellulären Ca2+ Konzentration im postsynaptischen Neuron des IC. Diese modulatorische Wirkung von Acetylcholin könnten damit eine wichtige Rolle bei der Entwicklung und Stabilisierung von inhibitorischen Synapsen im sich entwickelnden IC der Ratte spielen.
Die Hitzestressantwort stellt einen universellen Schutzmechanismus aller lebenden Organismen dar. Infolge einer Temperaturerhöhung werden Hitzestresstranskriptionsfaktoren (Hsf) aktiviert und bewirken eine gesteigerte Expression von Hitzestressproteinen (Hsp). Als molekulare Chaperone schützen diese die Zelle vor durch Hitze verursachten Schäden. In höheren Pflanzen ist dieses Phänomen sowohl auf der Ebene der Hsf als auch der Hsp besonders komplex. Das Ziel der vorliegenden Arbeit war die Untersuchung der Funktion von Komponenten des Chaperonsystems in der pflanzlichen Thermotoleranz. Zur Untersuchung der Thermotoleranz wurde ein transientes Expressionsystem mit Mesophyllprotoplasten aus steril angezogenen Tomatenpflanzen (Lycopersicon esculentum) zweier Linien (WT und CS) verwendet. CS-Pflanzen zeigen Cosuppression von HsfA1 und zeichnen sich durch eine Integration zweier direkt aufeinander folgender Transgenkassetten in invertierter Orientierung aus. Die fehlende Expression von HsfA1 in CS-Pflanzen ist die Folge eines Prozesses, der als RNA-interference (RNAi) bezeichnet wird. In unserem transienten Expressionssystem wurden Mesophyllprotoplasten mit einem Expressionsplasmid transformiert, das für Luciferase aus Photinus pyralis als thermosensitivem, leicht nachweisbarem Reporterprotein kodiert. Mit Hilfe dieses Testsystems konnten wir den Schutz der Luciferase gegen eine thermische Denaturierung bei 41°C (30 min) und die nachfolgende Renaturierung für 120 min bei 25°C in Abhängigkeit von endogenen und transient exprimierten Hsp und Hsf beobachten. Mit Hilfe der RNAi-Technologie und unter Verwendung von genspezifischen inverted repeat-Konstrukten konnten wir weiterhin die Bildung einzelner Komponenten des endogenen Chaperonsystems verhindern und damit ihre Funktion untersuchen. Es zeigte sich, dass in Protoplasten aus CS-Pflanzen praktisch alle hitzestressinduzierten Proteine fehlten und diese nicht in der Lage waren, Thermotoleranz auszuprägen, wie unter Verwendung des Reporterproteins Luciferase nachgewiesen werden konnte. Weiterhin fand keine Bildung cytoplasmatischer Multichaperonkomplexe, der sogenannten Hitzestressgranula (HSG), statt. Dieser Defekt in der Ausprägung von Thermotoleranz konnte durch Expression von HsfA2, HsfA3 und HsfA4b repariert werden. Die Überexpression dieser Hsf führte gleichermaßen zu (1) einer Expression von Chaperonen, (2) Thermoprotektion des Reporterenzyms Photinus pyralis-Luciferase und (3) Bildung von HSG-Komplexen. In weiteren Analysen lag unser Augenmerk insbesondere auf Vertretern der sHsp, sowie der Hsp70- und Hsp101-Chaperonfamilien. Hierbei erwies sich, dass vor allem Klasse CI-sHsp und Vertreter der Hsp70-Famile beim Schutz der Luciferase gegen Denaturierung während eines Hitzstresses eine Rolle spielen, während hauptsächlich Hsp101 und Vertreter der Hsp70-Familie in der darauf folgenden Erholungsphase von Bedeutung sind. Die Untersuchung der Interaktionen von drei Klassen cytoplasmatischer sHsp und ihrer intrazellulären Verteilung im Rahmen meiner Arbeit zeigte, dass jeder dieser Klassen eine unterschiedliche Funktion im Netzwerk cytoplasmatischer sHsp zukommt. Unter Verwendung nativer Gelelektrophorese und indirekter Immunfluoreszenz konnte nachgewiesen werden, dass sHsp der Klassen CI, CII und CIII in der Lage sind, auf der Ebene oligomerer Komplexe zu interagieren und ihre intrazelluläre Lokalisation wechselseitig zu beeinflussen. Proteine der Klasse CII zeigten eine starke Tendenz zur Bildung von Aggregaten, in die Klasse CIII-sHsp rekrutiert wurden. Im Unterschied dazu verfügten Klasse CI-Proteine über die Fähigkeit, diese Aggregate aufzulösen. Die detaillierte Untersuchung von fünf Isoformen der Klasse CI und zwei Isoformen der Klasse CII aus Lycopersicon esculentum ergab, dass diese oligomere Komplexe einer unterschiedlichen Anzahl von Untereinheiten bilden. Nach Coexpression waren Proteine beider Klassen in heterooligomeren Komplexe zu finden. Allerdings deuteten sich bei der Analyse der Fähigkeit einzelner Isoformen der Klasse CI, Heterooligomere mit Klasse CII-Proteinen zu bilden, Unterschiede an. sHsp kommt weiterhin eine Funktion in der Kontrolle der Aktivität von HsfA2 zu. Im Rahmen dieser Arbeit konnte ich zeigen, dass sHsps der Klassen CI und CII völlig unterschiedliche Rollen in der Regulation der intrazellulären Verteilung von HsfA2 spielen. Nach Überexpression in Mesophyllprotoplasten bildete LpHsp17.4-CII, nicht aber das nahe verwandte LpHsp17.3-CII mit HsfA2 große, cytoplasmatische Aggregate. Hsp17-CI dagegen verhinderte die Coaggregation von Hsp17.4-CII mit HsfA2.
Here I analyse 23 populations of D. galeata, a large-lake cladoceran, distributed mainly across the Palaearctic. I detected high levels of clonal diversity and population differentiation using variation at six microsatellite loci across Europe. Most populations were characterised by deviations from H-W equilibrium and significant heterozygote deficiencies. Observed heterozygote deficiencies might be a consequence of simultaneous hatching of individuals produced during different times of the year or of the coexistence of ecologically and genetically differentiated subpopulations. A significant isolation by distance was only found over large geographic distances (> 700 km). This pattern is mainly due to the high genetic differentiation among neighbouring populations. My results suggest that historic populations of Daphnia were once interconnected by gene flow but current populations are now largely isolated. Thus local ecological conditions which determine the level of biparental sexual reproduction and local adaptation are the main factors mediating population structure of D. galeata. The population genetic structure and diversity in D. galeata was investigated at a European scale using six microsatellite loci and 12S rDNA sequence data to infer and compare historical and contemporary patterns of gene flow. D. galeata has the potential for long-distance dispersal via ephippial resting eggs by wind and other dispersing vectors (waterfowl), but shows in general strong population differentiation even among neighbouring populations. A total of 427 individuals were analysed for microsatellite and 85 individuals for mitochondrial (mtDNA) sequence data from 12 populations across Europe. I detected genetic differentiation among populations across Europe and locations within sampling regions for both genetic marker systems (average values: mtDNA FST = 0.574; microsatellite FST = 0.389), resulting in a lack of isolation by distance. Furthermore, several microsatellite alleles and one haplotype were shared across populations. Partitioning of molecular variance was inconsistant for both marker systems. Microsatellite variation was higher within than among populations, whereas mtDNA data yielded an inverse pattern. Relative high levels of nuclear DNA diversity were found across Europe. The amount of mitochondrial diversity was low in Spain, Hungary and Denmark. Gene flow analysis at a European scale did not reveal typical pattern of population recolonization in the light of postglacial colonization hypotheses. Populations, which recently experienced an expansion or population-bottleneck were observed both in middle and northern Europe. Since these populations revealed high genetic diversity in both marker systems, I suggest these areas to represent postglacial zones of secondary contact among divergent lineages of D. galeata. In order to reveal the relationship between population genetic structure of D. galeata and the relative contribution of environmental factors, I used a statistical framework based on canonical correspondence analysis. Although I detected no single ecological gradient mediating the genetic differentiation in either lake regions, it is noteworthy that the same ecological factors were significantly correlated with intra- and interspecific genetic variation of D. galeata. For example, I found a relationship between genetic variation of D. galeata and differentiation with higher and lower trophic levels (phytoplankton, submerged macrophytes and fish) and a relationship between clonal variation and species diversity within Cladocera. Variance partitioning had only a minor contribution of each environmental category (abiotic, biomass/density and diversity) to genetic diversity of D. galeata, while the largest proportion of variation was explained by shared components. My work illustrates the important role of ecological differentiation and adaptation in structuring genetic variation, and it highlights the need for approaches incorporating a landscape context for population divergence.
Mitochondial NADH:ubiquinone oxidoreductase (complex I) the largest multiprotein enzyme of the respiratory chain, catalyses the transfer of two electrons from NADH to ubiquinone, coupled to the translocation of four protons across the membrane. In addition to the 14 strictly conserved central subunits it contains a variable number of accessory subunits. At present, the best characterized enzyme is complex I from bovine heart with a molecular mass of about 980 kDa and 32 accessory proteins. In this study, the subunit composition of mitochondrial complex I from the aerobic yeast Y. lipolytica has been analysed by a combination of proteomic and genomic approaches. The sequences of 37 complex I subunits were identified. The sum of their individual molecular masses (about 930 kDa) was consistent with the native molecular weight of approximately 900 kDa for Y. lipolytica complex I obtained by BN-PAGE. A genomic analysis with Y. lipolytica and other eukaryotic databases to search for homologues of complex I subunits revealed 31 conserved proteins among the examined species. A novel protein named “X” was found in purified Y. lipolytica complex I by MALDI-MS. This protein exhibits homology to the thiosulfate sulfurtransferase enzyme referred to as rhodanese. The finding of a rhodanese-like protein in isolated complex I of Y. lipolytica allows to assume a special regulatory mechanism of complex I activity through control of the status of its iron-sulfur clusters. The second part of this study was aimed at investigating the possible role of one of these extra subunits, 39 kDa (NUEM) subunit which is related to the SDRs-enzyme family. The members of this family function in different redox and isomerization reactions and contain a conserved NAD(P)H-binding site. It was proposed that the 39 kDa subunit may be involved in a biosynthetic pathway, but the role of this subunit in complex I is unknown. In contrast to the situation in N. crassa, deletion of the 39 kDa encoding gene in Y. lipolytica led to the absence of fully assembled complex I. This result might indicate a different pathway of complex I assembly in both organisms. Several site-directed mutations were generated in the nucleotide binding motif. These had either no effect on enzyme activity and NADPH binding, or prevented complex I assembly. Mutations of arginine-65 that is located at the end of the second b-strand and responsible for selective interaction with the 2’-phosphate group of NADPH retained complex I activity in mitochondrial membranes but the affinity for the cofactor was markedly decreased. Purification of complex I from mutants resulted in decrease or loss of ubiquinone reductase activity. It is very likely that replacement of R65 not only led to a decrease in affinity for NADPH but also caused instability of the enzyme due to steric changes in the 39 kDa subunit. These data indicate that NADPH bound to the 39 kDa subunit (NUEM) is not essential for complex I activity, but probably involved in complex I assembly in Y. lipolytica.
The thesis entitled „Investigations on the significance of nucleo-cytoplasmic transport for the biological function of cellular proteins" aimed to unreveal molecular mechanisms in order to improve our understanding of the impact of nucleo-cytoplasmic transport on cellular functions. Within the scope of this work, it could be shown that regulated nucleo-cytoplasmic transport of a subfamily of homeobox transcription factors controlled their intra- and intercellular transport, and thereby influencing also their transcriptional activity. This study describes a novel regulatory mechanism, which could in general play an important role for the ordered differentiation of complex organisms. Besides cis-active transport Signals, also post-translational modifications can influence the localization and biological activity of proteins in trans. In addition to the known impact of phosphorylation on the transport and activity of STAT1, experimental evidence was provided demonstrating that acetylation affected the interaction of STAT1 with NF-kB p65, and subsequently modulated the expression of apoptosis-inducing NF-kB target genes. The impact of nucleo-cytoplasmic transport on the regulation of apoptosis was underlined by showing that the evolutionary conservation of a NES within the anti-apoptotic protein survivin plays an essential role for its dual function in the inhibition of apoptosis and ordered cell division. Since survivin is considered a bona fide cancer therapy target, these results strongly encourage future work to identify molecular decoys that specifically inhibit the nuclear export of survivin as novel therapeutics. In order to further dissect the regulation of nuclear transport and to efficiently identify transport inhibitors, cell-based assays are urgently required. Therefore, the cellular assay Systems developed in this work may not only serve to identify synthetic nuclear export and Import inhibitors but may also be applied in systematic RNAi-screening approaches to identify novel components of the transport machinery. In addition, the translocation based protease- and protein-interaction biosensors can be applied in various biological Systems, in particular to identify protein-protein interaction inhibitors of cancer relevant proteins. In summary, this work does not only underline the general significance of nucleo-cytoplasmic transport for cell biology, but also demonstrates its potential for the development of novel therapies against diseases like cancer and viral infections.
Calcium-activated potassium channels are fundamental regulators of neuron excitability. SK channels are activated by an intracellular increase of Ca++ (such as occurs during an action potential). They have a small single channel conductance (less than 20pS) and show no voltage dependence of activation. To date, there are only a few examples of high-resolution structures of eukaryotic membrane proteins. All of them were purified from natural sources. Since no abundant natural sources of eukaryotic K+ channels are available we overexpressed rSK2 in order to produce the quantities necessary for structural analysis. Unfortunately the Pichia pastoris expression system did not yield sufficient amount of pure protein, mainly because most of the protein was retained by in the ER and was only partially soluble. Subsequently, two constructs were expressed: SK2-FCYENE (containing a specific sequence that promotes surface expression), and SK2-q-CaM a concatamer of SK2 and calmodulin. Although these proved an improvement in terms of solubilisation, little improvement was found in terms of amounts of purified material obtained. For this reason we tested the Semliki Forest virus expression system, since the protein is expressed in a mammalian system where we hoped that it would be trafficked in the same way as in vivo. Using this system it was possible to express rSK2 and solubilise it with several detergents and to achieve much better purification. However, the levels were still not sufficient for high-resolution structural studies, although sufficient for single particle electron microscopy analysis.
Holzkohlen aus archäologischen Grabungen im Sahel von Burkina Faso belegen die regionale Geschichte der Gehölzvegetation über die letzten 2000 Jahre. Der Bodenbau, den die sesshaft lebende Bevölkerung seit Beginn unserer Zeitrechnung intensiv betreibt, veränderte die Zusammensetzung der Gehölzvegetation vor allem auf den Dünen der Region. Im Vergleich mit der heutigen Vegetation lassen sich zudem klimatische Veränderungen nachweisen. Untersucht wurden über 9000 Fragmente aus sieben verschiedenen archäologischen Grabungen. Sechs Inventare stammen aus Siedlungshügeln. Bei einem Fundplatz handelt es sich um einen Hausgrundriss. Insgesamt wurden 37 Holzkohletypen erkannt und dokumentiert. Die untersuchten Inventare der Siedlungshügel zeigen, dass vor allem die Gehölzvegetation der Dünen und der Galeriewälder zur Brennholzentnahme genutzt wurde. Je nach Lage der Siedlung und dem Schwerpunkt der Wirtschaftsweise können verschiedene Taxa mit höheren Anteilen vertreten sein, möglicherweise zusätzlich verstärkt durch die anthropogene Auswahl von verfügbarem Brennholz. Im Vergleich der Holzkohleinventare lassen sich für die Eisenzeit regionale Entwicklungen erkennen. Die natürlichen Gehölzbestände auf den Dünen, unter anderem aus verschiedenen Akazienarten, wurden, zumindest in der Umgebung der Siedlungen, verdrängt. Stattdessen nahmen aufgrund der selektiven Förderung durch den Menschen die Anteile der Gehölze der Kulturbaumparks, Vitellaria paradoxa und Faidherbia albida zu. Die Landwechselwirtschaft förderte zudem Brachearten insbesondere aus der Familie der Combretaceae. In der späten Eisenzeit nahm Guiera senegalensis zu, die von starker Beweidung der Brachen profitiert. Der Unterwuchs der Galeriewälder an den mares und Wasserläufen wurde mit zunehmender Besiedlungsdauer in der Umgebung der einzelnen Fundplätze aufgelichtet, die Anteile von Combretum micranthum gehen in den Inventaren der einzelnen Siedlungsplätzen jeweils zurück. Klima und Vegetation waren während der Eisenzeit sudano-sahelisch. Auf feuchteres Klima verweisen Vitellaria paradoxa und Detarium microcarpum, die deutlich höhere Niederschläge benötigen, als sie die Region heute erhält. Der hohe Anteil von Taxa, die heute weiter südlich verbreitet sind, belegt zudem den sudanischen Aspekt der Gehölzvegetation. Der Vergleich der anthrakologischen mit den palynologischen und karpologischen Ergebnissen zeigt, dass die Gehölzvegetation sich unter zunehmend arideren Bedingungen in den letzen 2000 Jahren anthropozoogen stark verändert hat. Das Klima scheint aber während der Eisenzeit von 0-1500 AD vergleichsweise stabil gewesen zu sein. Erst danach haben die Niederschläge sich soweit verringert, dass in den letzten 500 Jahren einige sudanische Taxa aus der Region verschwanden, die noch während der Eisenzeit zur regionalen Flora gehört hatten, zum Beispiel Vitellaria paradoxa, Detarium microcarpum und Lannea sp. Der Vergleich der eisenzeitlichen Holzkohleflora mit der rezenten Dynamik der Vegetation und mit der Verbreitung einiger Arten um die Mitte des 20. Jahrhunderts zeigt, dass einige Taxa, wie Terminalia sp. möglicherweise erst in den letzten fünfzig Jahren aus der Region verschwunden sind.
Die Medusen der metagenetischen Cnidarier (Nesseltiere) werden je nach Kultur- und Sprachraum z. B. mit einer weiblichen Gestalt mit schlangenartigen Haaren (Medusa), mit stark brennenden Quaddeln auf der Haut oder mit Stränden verklebenden Gallertmassen in Verbindung gebracht. Diese Assoziationen fußen hauptsächlich auf Merkmalen der Scheibenquallen (Scyphozoa), deren bisexuelle Medusen in Größe, Tentakellängen, Nematocytenbesatz und Mesogloeagehalt gegenüber der relativ unauffälligen ungeschlechtlichen Polypengeneration beeindrucken. Häufig ist die Medusengeneration der Scheibenquallen namensgebend. Die Medusen der Ohrenqualle Aurelia aurita (Fahnenquallen, Semaeostomeae) sind an den durchscheinenden, ohrenförmig um den Gastralraum angeordneten Gonaden erkennbar. In gemäßigten Klimazonen ist die bisexuelle Vermehrung - und das anschließende oft massenhafte Absterben in Küstennähe - auf den Sommer beschränkt (Lucas 1996), so daß die Medusen der Ohrenqualle häufige spätsommerliche Gäste an Stränden z. B. der Nord- und Ostsee (mit für den Menschen harmloser Nesselwirkung) sind. Vertreter der von Linné (1758) und Lamarck (1816) typologisierten Aurelia aurita sind weltweit verbreitet und bezüglich der Morphologie, Physiologie, Ethologie und Ökologie gut untersucht (z. B. Fautin & Lowenstein 1992; Costello & Colin 1994; Hammer et al. 1994; Lucas 1994). Unter ökologischen Gesichtspunkten ist das breite Spektrum der Ohrenqualle bezüglich abiotischer Umweltfaktoren hervorzuheben. Die häufig massenhaften Medusenaggregationen und der daraus resultierende Predationsdruck gegenüber dem Mikrozooplankton (Copepoden, Invertebratenund Fischlarven) weist der Ohrenqualle eine wichtige - und gelegentlich dominierende - Stellung in marinen Plankton-Lebensgemeinschaften zu (Janas & Witek 1993; Schneider & Behrends 1994; Sullivan et al. 1994; Tsikhon-Lukanina et al. 1996). Die breite Anpassungsfähigkeit und hohe Vermehrungsrate wird anekdotenhaft durch eine kürzlich erfolgte Meldung bekräftigt, nach der offensichtlich Aurelia-Schwärme die Kühlwasserleitungen eines Kraftwerks in Australien verstopften und lahmlegten (Cnidaria-List-Server). Das ubiquitäre Vorkommen läßt sich auf eine große ökologische Toleranz gegenüber Klima und Salzgehalt zurückführen, so daß die Ohrenqualle als Bewohner aller Meere von 40° südlicher bis 70° nördlicher Breite und Habitaten mit Salinitäten von 6-41‰ gilt (Kramp 1961). Dabei werden geographische Populationen, Unterarten und Arten der Gattung Aurelia anhand morphologischer Medusenmerkmale sowie auf Basis von Allozymunterschieden in unterschiedlicher Konsequenz teilweise in eine Art, Aurelia aurita zusammengefaßt, oder in mehrere distinkte Spezies voneinander abgegrenzt (Mayer 1910; Kramp 1961; Russell 1970; Kozloff 1987; Greenberg et al. 1996). Der nicht konsistente taxonomische Status der Ohrenqualle beruht entweder auf phylogenetisch uninformativen diagnostischen Merkmalen oder spiegelt Differenzierungen wider, die mit Artbildungsprozessen erklärt werden können. In der vorliegenden Arbeit sollen die phylogenetischen Beziehungen zwischen Aurelia-Populationen auf einer weltweiten geographischen Skala unter organismischen und molekularen Aspekten untersucht werden. Die möglichen Speziationsprozesse orientieren sich an den von Avise (2000) klassifizierten beiden Kategorien, nach denen die gängigen Spezieskonzepte auf Basis phylogenetischer und biologischer Kriterien, oder analog von Ridley (1996) nach evolutionären und zeitlich unabhängigen Kriterien eingeteilt werden. Die DNA-Sequenzinformationen eines mitochondrialen Gens (16S rDNA) sowie eines auf dem Kerngenom liegenden ribosomalen Locus (ITS-1/5.8S rDNA) sollen verwendet werden, um die genealogischen Beziehungen verschiedener Aurelia- Populationen sowohl phylogeographisch (Avise 1994) als auch bezüglich der phylogenetischen Artkonzepte zu untersuchen (Eldredge & Cracraft 1980; Wiley 1981; Cracraft 1989; Übersicht in Hull 1997). Mittels morphologischer Merkmale, Lebenszyklusdaten und den molekulargenetischen Daten soll gleichzeitig geprüft werden, ob sich die von Avise (2000) genannte Kategorie der biologisch definierten Spezieskonzepte in phänetisch (Sneath & Sokal 1973; Whittemore 1993), ökologisch (van Valen 1976; Schluter 1996) oder reproduktiv (Mayr 1963) getrennte Einheiten bei der Ohrenqualle widerspiegeln. Der Beitrag verschiedener Diversifizierungsfaktoren wie geographische Verbreitung, Selektion und genetische Drift wird in diesem Zusammenhang unter besonderer Berücksichtigung ökologischer Aspekte in Kapitel 2 dargestellt. Nicht zuletzt auch aufgrund ihrer besonderen ökologischen Plastizität empfiehlt sich die Ohrenqualle zur Nutzung als Biomonitor zur Untersuchung von Umweltbeeinträchtigungen auf Individuumebene. Eine in diesem Zusammenhang nützliche Besonderheit im Lebenszyklus von Aurelia betrifft den Übergang von der sich ungeschlechtlich vermehrenden Polypengeneration in die bisexuelle Medusengeneration, bei dem an der Oralseite des Polypen Medusenlarven (Ephyren) in Form einer polydisken Strobilation abgeschieden werden. Die Strobilation wird extrinsisch durch natürliche Faktoren ausgelöst und kann bei den Polypen der Ohrenqualle künstlich durch Temperaturabsenkung oder Erhöhung der Iod-Ionenkonzentration induziert werden (Spangenberg 1967). Dieses experimentell beeinflußbare Lebenszyklusstadium wurde im Rahmen von ökotoxikologischen Studien verwendet, um den Einfluß von Chemikalien auf die verzögerte Strobilation nach künstlicher Auslösung oder die Störung des Strobilationsverlaufs zu testen (Spangenberg 1984; Thiel & Jarms 1986). Black & Bloom (1984) untersuchten die Beziehung zwischen der Strobilationsinduktion und der Produktion von Hitzeschockproteinen (HSP) nach Temperaturerhöhung und konnten die Induzierbarkeit von Hitzeschockproteinen als Reaktion auf Temperaturstreß immunologisch nachweisen. Diese Studien zeigen, daß die Ohrenqualle ein hohes Reaktionspotential gegenüber variablen Umweltbedingungen besitzt. Die Charakterisierung dieses Reaktionspotentials soll in der vorliegenden Arbeit mittels der Analyse organismischer Reaktionen gegenüber anthropogen verursachten Streßfaktoren durchgeführt werden. Im Rahmen von experimentellen Schadstoff- expositionen sollen an Aurelia-Polypen genetische Reaktionen auf der Ebene der mRNATranskription untersucht werden. Der Schwerpunkt liegt auf der Etablierung von Biomarkern (van Gestel & van Brummelen 1996) mittels molekulargenetischer Methoden, wobei die Durchführbarkeit und Effektivität verschiedener Methodenansätze verglichen werden sollen. Die identifizierten Biomarker sollen im Hinblick auf die Analyse der Dosis-Wirkungsbeziehungen zwischen Schadstoffkonzentrationen und dem Ausmaß der genetischen Reaktion mittels quantitativer PCR-Verfahren näher charakterisiert werden. Ein anschließender Test der im Labor charakterisierten Biomarker auf die Anwendbarkeit im Freiland soll entsprechende Hinweise für die Sensitivität und Durchführbarkeit des in Kapitel 3 ausgeführten Biomarker-Assays liefern. Ein weiteres Ziel der vorliegenden Arbeit ist die Verknüpfung verschiedener biologischer Organisationsebenen im Kontext ökologischer und ökotoxikologischer Forschung (Clements 2000). Erkenntnisse zum Wirken von Umwelteinflüssen auf molekularer Ebene bieten sich als Basis einer Bewertung individueller Reaktionen für Veränderungen auf Populationsebene an. Dieser Zusammenhang wird in Kapitel 4 anhand der Relevanz von Biomarkern für Aussagen auf Populationsund Speziesebene besprochen.
