G-protein coupled receptors (GPCRs) comprise the largest superfamily of cell surface receptors and possess a signature motif of seven transmembrane helices. The endothelin B (ETB) receptor is a member of rhodopsin like GPCR family. It plays an important role in vasodilation and is found in the membranes of the endothelial cells enveloping blood vessels. Knowledge of the three-dimensional structure of G-protein coupled receptors in general would significantly add to our understanding of their molecular mechanisms and would be useful in the search for new specific drugs. However, three-dimensional structural analysis will require milligram quantities of pure and homogeneous protein. This dissertation is a study of the production, biochemical characterization and preliminary structural studies of the human ETB G-protein coupled receptor. The present work aimed at elucidating the structure and mechanistic details of function of the receptor by using a combination of X-ray crystallographic and NMR methods for collecting structural data. To obtain homogenous and monodisperse receptor protein preparation for structural and functional studies, we implemented the baculovirus expression system for the production of ETB receptor for the present work. The two step affinity purification ensured capture of full-length receptor. Silver stained SDS-PAGE of the purified receptor-ligand complex indicated greater than 90% protein purity. Based on previous reports, we used the high affinity ligand (endothelin -1) binding to the receptor for co-crystallization of receptor-ligand complex by locking the receptor in the activated conformation. As a prerequisite for 3D crystallization trials, the stability of the detergent solubilized receptor-ligand complex was assessed with respect to pH, temperature and time. Receptor-ligand complex did not show any degradation and aggregation over 6 days at 4°C and 18°C. Interestingly, change of pH suggested that receptor-ligand complex is unstable at lower pH due to possible charge induced conformational changes. In our work, we introduced the idea of using fluorophore labeled ligand for simple visual recognition of the receptor-ligand complex during purification and crystallization. On the other hand, we alternatively used biotinylated endothelin-1 to produce an adequate amount of ligand bound receptor complex, thus ensuring homogeneity of the purified complex for use in structural studies. Thus far, preliminary crystals have been obtained for both the unlabelled ET-1 and fluorophore labeled ET-1 complexed with ETB receptor. Moreover, we performed the systematic investigation of the protein/peptide binding partner for the receptor-ligand complex with the chief aims of stabilizing structure and increasing the possibilities of 3D-crystal contacts. Thus subsequent to formation of receptor-ligand complex, the additional in vitro formation of a ternary arrestin-receptor-ligand complex was also attempted for use in structural studies. We successfully demonstrated that arrestin mutant (R169E) forms a tight complex with ETB receptor regardless of its phosphorylation state. A second approach to get insight into the ETB receptor ligand binding site relied on the use of spin isotope labeled ET-1 ligand peptide by employing solid state MAS NMR method. Preliminary data provided compelling evidence that the C-terminal region of the peptide is immobilized in an ordered environment and presumably bound to the receptor. This indicates that the approach is feasible, although there are difficulties in sample preparation for further spectral measurements and data collection which are currently being discussed in ongoing investigations. At this point of our research work, we initiated a collaborative effort to obtain high yields of pure, active receptor without post translational modifications, from an E. coli cell lysate based in vitro expression system. We successfully optimized the production of homogenous and monodisperse endothelin B receptor in mg amounts. Thus this could potentially provide an alternative source of high quality receptor production in large quantities for immediate crystallization trials. Thus we hope that the results from these investigations can be applied in a more general sense to the production and crystallization of other G protein-coupled receptors.
Die vorliegende Arbeit fasst folgende experimentellen Arbeiten zusammen: • Synthese von amorphem, schwarzem, basischem Silicium Si am,schw,ox und amorphem, schwarzem, nicht basischem Silicium Siam,schw durch Reduktionsreaktion von Siliciumtetrachlorid mit Natrium. • Reaktivität des amorphen Siliciums Si am gegenüber verschiedenen Gasen: Cl2, HCl, CH3Cl • Reaktivität von Si am gegenüber Alkoholen bei Raumtemperatur. • Reaktivität von Si am gegenüber Alkoholen und Essigsäure in einem geschlossenen System, nicht katalysiert/katalysiert durch Cu(I)Cl und Cu(I)O. • Reaktivität von Si am,schw,ox und Si am,schw gegenüber Alkoholen und Essigsäure in einem offenen System (Ofen, „slurry phase“-Reaktor), nicht katalysiert/katalysiert durch Cu(I)Cl und Cu(I)O. • Thermische und katalytische Disproportionierungsreaktionen von Methylmethoxysilanen, katalysiert durch Alkalimetalle (Na), salzartige Verbindungen (Ca(OH)2, MgSO4/C, Na2SO4/C und (NH4)2SO4/C) und Lewis-Säuren (AlCl3). • Komproportionierung zwischen Trimethylmethoxysilan und Methyltrimethoxysilan Der Schwerpunkt dieser Arbeit lag in Untersuchung der chemischen Reaktivität von amorphem Silicium Si am, synthetisiert durch Na-Reduktion von Siliciumtetrachlorid, gegenüber verschiedenen Gasen, wie z. B. Chlorgas, Chlorwasserstoff und Methylchlorid, gegenüber Alkoholen, wie z. B. Methanol, Ethanol und Phenol und gegenüber Essigsaure unter unterschiedlichen Reaktionsbedingungen. Die Reaktion zwischen amorphem Silicium Si am und Cl2-Gas führt bei 240-250°C zu Tetrachlorsilan SiCl4 als einziges Produkt. Die Reaktion mit HCl-Gas liefert im Temperaturbereich zwischen 360-370°C zwei Produkte: 15% Dichlorsilan, H2SiCl2 und 85% Trichlorsilan, HSiCl3. Im Temperaturbereich zwischen 370-420°C entstehen drei Produkte: 36,4% Dichlorsilan H2SiCl2, 58,5% Trichlorsilan HSiCl3 und 5,1% Tetrachlorsilan SiCl4. Über eine Temperaturführung kann die Produktbildung wesentlich beeinflusst und damit auch gesteuert werden. Durch eine Reaktion mit Methylchlorid bei 560°C entstehen zwei Produkte: 79% Methyltrichlorsilan CH3SiCl3 und 21% Dimetyldichlorsilan (CH3)2SiCl2. Basisches Silicium Siam,schw,ox liefert in den Reaktionen mit Alkoholen und Essigsäure unter Rückfluss-Bedingungen (Temperaturen zwischen 20 und 420°C; offene Reaktionssysteme) jeweils ein einziges Produkt und führt mit den korrespondierenden Partnern selektiv zu Tetramethoxy-, Tetraethoxy-, Tetraphenoxy- und Tetraacetoxysilan. Diese Reaktionen werden durch die im eingesetzten Siam/NaCl-Gemisch vorhandene Base (NaOH, Na2Ox; x = 1, 2) katalysiert. Gemischte Alkylalkoxysilane oder Siloxane entstehen unter den vorgegebenen Bedingungen nicht. Silicium setzt sich dabei in den Reaktionen mit Methanol und Ethanol vollständig um. Werden die Bedingungen modifiziert und die Reaktionen in geschlossenen Systemen (Reaktionsampullen) bei 150°C, katalysiert durch 5 Gew. % Cu(I)Cl im Mol-Verhältnis Si am,schw,ox/CH3OH 1:3 und 1:4 durchgeführt, wird Trimethoxysilan in 91% und 84% Ausbeute gewonnen. Der Produktbildungsweg führt offensichtlich über die Katalyse eines Komplexes Na+[Cu(OH)Cl]-, der in situ aus Cu(I)Cl und NaOH gebildet wird. Nicht basisches Silicium, Siam,schw, reagiert mit Methanol bei Raumtemperatur zu Trimethoxysilan HSi(OCH3)3 (16-18%) und zu Tetramethoxysilan Si(OCH3)4 (84-82%). Die Produktbildung kann durch Änderungen Reaktionsbedingungen in Richtung von Trimethoxysilan verschoben werden. So entsteht Trimethoxysilan in einer geschlossenen Ampulle zu 95% als Hauptprodukt der Reaktion zwischen amorphem Silicium und Methanol im Mol-Verhältnis 1:3 in Anwesenheit von NH4HF2 als Aktivator. 84,5% Trimethoxysilan entstehen in einer Ampulle bei 200°C nach 5 Stunden Reaktionszeit, dann wenn Methanol im stöchiometrischen Verhältnis 8:1 eingesetzt wird. Der Umsatz an Silicium ist praktisch vollständig (100%). Wird die Reaktion zwischen Siam,schw und Methanol in einem „slurry phase“-Reaktor durchgeführt, resultiert eine Abhängigkeit der Produktausbeute vor allem vom Lösungsmittel, dann aber auch von den weiteren Bedingungen. In Dodecylbenzol entsteht das Trimethoxysilan bei 230°C und mit Cu(I)O als Katalysator zu 72% in einer 8-stündigen Reaktion. Unter vergleichbaren Bedingungen, aber in Isoparaffinöl bildet sich Trimethoxysilan zu 61%. In einem modifizierten „slurry phase“-Reaktor, in dem die Produkte mit überschüssigem Methanol sofort aus der Reaktion abgeführt werden, entsteht das Trimethoxysilan nicht. Allerdings bilden sich in diesem Fall methylierte Methoxysilane. Der Umsatz an Silicium hängt auch von der Temperatur des Methanol-Dampfes ab. Wird die Temperatur zwischen 150-200°C gehalten, werden 49% des Siliciums umgesetzt. Ansonsten beträgt die Umsatzrate des Siliciums 20-25%. Amorphes, schwarzes, basenfreies Silicium, Si am,schw, reagiert bei 150°C mit Ethanol im Mol-Verhältnis 1:3 zu 91% und bei 200°C (Mol-Verhältnis Si : EtOH 1:8) zu 86% Triethoxysilan, HSi(OEt)3. Die Reaktion zwischen nicht basischem Silicium Si am,schw und Essigsäure liefert in der Siedehitze kein Tetraacethoxasilan, sondern nur Polyacethoxysilane mit einem Polymerisationsgrad von n = 2, 3, in einer Ampulle bildet sich jedoch ein Gemisch aus Tetraacetoxysilan und verschiedenen Acethoxypolysilanen. Die Polysilanbildung verstärkt sich mit zunehmender Reaktionstemperatur und –dauer. Die Reaktion zwischen Si am,schw und Essigsäure in einem offenen System (Ofen) liefert bei 300°C außer Tetraacetoxysilan ein Gemisch von Acethoxypolysilanen mit n=2-6. Versuche zur Bildung von Phenoxsilanen aus Si am,schw. und Phenol schlugen bei Temperaturen zwischen 150-300°C fehl. Das MALDI-TOF-MS-Spektrum einer bei 225-230°C im Vakuum siedenden Fraktion, die aus Reaktion von Si am,schw mit Phenol bei 420°C im Ofen entstanden ist, zeigt Spuren von Tetraphenoxysilan und organische Polymere mit dem Molekulargewichht bis zu 960 Dalton. Während Umsetzungen von amorphem Silicium mit Methanol in geschlossenen Reaktionsampullen Trimethoxysilan und Tetramethoxysilan als Hauptprodukte liefern, bilden sich in einem offenen System methylierte Methoxysilane MenSi(OMe)4-n (n=1, 2) in unterschiedlichen Ausbeuten. Dimethyldimethoxysilan entsteht aus basischem Silicium Siam,schw,ox nicht; selbst durch Katalysatorzusatz entsteht das gewünschte Produkt nur in Spuren. Die Monomethylierung verläuft dagegen erfolgreicher. Die höchste Ausbeute an Methyltrimethoxysilan (33,7%) wird bei 320°C mit 20 Gew. % Cu(I)Cl als Katalysator erzielt. Völlig anders verhält sich nicht basisches Silicium Si am,schw , denn es reagiert mit Methanol in einem offenen System bei 350°C zu 13% Dimethyldimethoxysilan (CH3)2Si(OCH3)2, und 37% Methyltrimethoxysilan CH3Si(OCH3)3. In Reaktionen mit größeren Silicium-Mengen (Si-Gehalt: 39-60 mmol) entstehen in einem nicht gerührten Reaktionsreaktor nach 20’ Minuten Reaktionszeit bei 300°C 26% Dimethyldimethoxysilan (CH3)2Si(OCH3)2 und nach 45’ Minuten Reaktionszeit 49,5% Methyltrimethoxysilan CH3Si(OCH3)3. In einem nicht gerührten Reaktionsrohr entstehen nach 3 Stunden Reaktionszeit 25-30% Dimethyldimethoxysilan bei 300°C und mit 20 Gew. % Kupfer(I)Chlorid als Katalysator. In einem gerührten Reaktionsrohr bilden sich bei 350°C und mit 20 Gew. % Cu(I)Cl nach 3-stündiger Reaktion 23,5% (CH3)2Si(OCH3)2 und 53,3% CH3Si(OCH3)3. Das Vorhandensein des Katalysators erhöht die Ausbeute an methylierten Methoxysilanen in den Reaktionen zwischen nicht basischem Silicium und Methanol. Aus dem Verlauf der Reaktion im Ofen lässt sich schließen, dass die größte Menge des Dimethyldimethoxysilans, (CH3)2Si(OCH3)2, während der ersten 20-40 Minuten entsteht. Danach sinkt der Anteil mit zunehmender Reaktionszeit, es bilden sich verstärkt die festen Dimethyloligosiloxane. Die Ausbeute an Tetramethoxysilan, Si(OCH3)4, wächst mit der Reaktionszeit kontinuierlich. Dagegen bleibt die Ausbeute an Methyltrimethoxysilan, CH3Si(OCH3)3, während 3-stündiger Reaktion relativ konstant. Die Bildung aller drei Produkte wurde zwischen 300° und 350°C detailliert verfolgt. Nur ca. 20% des Siliciums setzen sich zu den flüchtigen Produkten um, rund 80% Silicium bleiben in fester Form als nicht abreagiertes Silicium und als feste Oligodimethylsiloxane erhalten. Eine nicht katalysierte Reaktion zwischen Si am,schw und Ethanol liefert nach 3h Reaktionszeit in einem nicht rührenden Reaktor etwa 23% HSi(OCH3)3 und etwa 72% Si(OEt)4. Etylenethoxysilane oder -siloxsane bilden sich nicht. Zum Weiteren ist es gelungen, Dimethyldimethoxysilan durch Disproportionierungsreaktion aus Methyltrimethoxysilan und Tetramethoxysilan über metallischem Natrium im Mol-Verhältnis 1:1 in einer Ampulle bei 250°C herzustellen. Aus Disproportionierung von Methyltrimethoxysilan sind 12% Dimethyldimethoxysilan zu erhalten. Die Disproportionierung von Tetramethoxysilan führt zu 27% Dimethyldimethoxysilan, (CH3)2Si(OCH3)2, 34% Methyltrimethoxysilan, CH3Si(OCH3)3, und 11% Trimethylmethoxysilan, (CH3)3SiOCH3. Die Menge des eingesetzten Natriums muss in weiteren Arbeiten noch optimiert werden. Eine Komproportionierungsreaktion zwischen Trimethylmethoxysilan, (CH3)3SiOCH3, und Methyltrimethoxysilan, CH3Si(OCH3)3, zu Dimethyldimethoxysilan, (CH3)2Si(OCH3)2, fand unter den in dieser Arbeit beschriebenen Bedingungen nicht statt. Diese Ergebnisse sind ein im Labormaßstab chlorfreies Verfahren zur Herstellung von methylierten Methoxysilanen. Unter der Voranstellung der Aufskalierbarkeit wird damit eine neue Route eines möglicherweise technisches Prozesses (das Q-Verfahren) zu Darstellung von Siliconen zugänglich; diese führt wie folgt aus: Siliciumtetrachlorid → amorphes Silicium → Tetramethoxysilan/Tetraethoxysilan/Tetraacetoxysilan → Methylmethoxysilane → Silicone. Es gilt nun, die Synthesebedingungen nach erfolgreicher Optimierung in den Pilotmaßstab zu überführen. Die Synthese von Trimethoxysilan, HSi(OCH3)3, erscheint bereits jetzt schon den Bedingungen des technisch durchführenden Cromptonprozesses überlegt zu sein. Diese bietet jedoch den Vorteil, von technisch verfügbarem Silicium anzugehen. Eine exakte und vergleichende Prozess-Analyse wird darüber Aufschluss geben, ob die in dieser Arbeit erworbenen Ergebnisse zu einer technischen Umsetzung führen.
Ferrocenbasierte Polymere haben in den vergangenen Jahrzehnten reges Interesse hervorgerufen, welches in ihren einzigartigen optischen und elektronischen Eigenschaften begründet ist. Diese sind auf die Anwesenheit der redoxaktiven Eisenionen sowie auf kooperative Effekte entlang des Polymerstrangs zurück zu führen. Die substanzspezifischen Merkmale hängen dabei nicht nur von der Position der Ferroceneinheiten im Makromolekül ab, sondern auch von der chemischen Natur der verbrückenden Einheiten. Hier bewähren sich solche Atome und Gruppen, die in der Lage sind, Elektronendichte zu übertragen. Ein in dieser Hinsicht besonders viel versprechendes Element ist Bor, da es im sp2-hybridisierten Zustand ein leeres p-Orbital besitzt und dieses für die Übertragung von Ladungsdichte zur Verfügung stellen kann. Vor diesem Hintergrund galt es nun auf der Basis neuartiger Kondensationsreaktionen zunächst Diferrocenylborane zu synthetisieren und anhand ausgewählter Derivate Kenntnisse über charakteristische Eigenschaften dieser Substanzklasse zu gewinnen. Die kurzkettigen Moleküle dienten als Modellverbindungen für Poly(ferrocenylen)e, welche im Mittelpunkt des zweiten Teils dieser Arbeit standen. .... In der vorliegenden Arbeit wurde gezeigt, dass sich das neuartige Polymer [-fc-B(Br)-]n (33) hervorragend als Edukt eignet, um vielfältige borverbrückte Poly(ferrocenylen)-Derivate sowohl mit trigonalen als auch mit tetragonalen Borzentren darzustellen. Die beschriebenen Systeme weisen eindeutige Merkmale von Eisen-Eisen-Wechselwirkungen entlang der Polymerhauptkette auf, wobei sich das Ausmaß dieser Interaktionen über die Wahl der Substituenten an den Boratomen gezielt beeinflussen lässt. Die dargestellten Materialien besitzen somit großes Potential für die Entwicklung „Molekularer Drähte“ auf der Nanoskala.
Bis zur Entfaltung der Wirkung eines Pharmakons laufen zahlreiche komplexe Vorgänge ab, die sich in drei wesentliche Phasen unterteilen lassen. Die pharmazeutische Phase umfasst mit der Applikation und dem Zerfall der Arzneiform sowie dem Auflösen des Wirkstoffes Vorgänge, die im wesentlichen von den galenischen Eigenschaften des Arzneistoffes abhängen. In der pharmakokinetischen Phase erfolgt mit der Resorption die Aufnahme des Wirkstoffes in den Organismus, dem sich die Verteilung in die unterschiedlichen Gewebe über die Blutbahn anschließt. Durch verschiedene Eliminationsprozesse wird der Wirkstoff zuletzt wieder aus dem Körper ausgeschieden. Mit dem Erreichen des Wirkortes beginnt die pharmakodynamische Phase, in der die pharmakologischen Effekte des Arzneistoffes zur erwünschten klinischen Wirkung führen. Der Nachweis des Pharmakons am Wirkort in klinisch-relevanten Konzentrationen ermöglicht somit Rückschlüsse auf die Wirksamkeit des Arzneistoffes, aber unter bestimmten Voraussetzungen auch auf dessen Wirkmechanismus. Während mittlerweile ein Großteil neuer Arzneistoffe mit Hilfe unterschiedlicher Mechanismen durch exaktes Drug Targeting an den Wirkort gesteuert werden, weisen andere Substanzen teilweise ungewollt eine gewebespezifische, dirigierende Komponente auf, die neben der eigentlichen Hauptwirkung weitere unterstützende oder auch unerwünschte Effekte auslösen. Von besonderem Interesse ist diese Gewebespezifität für pflanzliche Arzneistoffe, deren Wirkkomponenten bislang noch nicht eindeutig bestimmt werden konnten. Gemeinsam mit anderen pharmakologischen Befunden kann der analytische Nachweis eines wirksamkeitsmitbestimmenden Inhaltsstoffes am Wirkort in ausreichenden Konzentrationen ein weiterer deutlicher Hinweis auf dessen Wirkbeteiligung sein. Besonders vor dem Hintergrund einer rationalen, evidenz-basierten Pharmakotherapie, deren Anforderungen die pflanzlichen Arzneien mittlerweile ebenso wie die synthetischen Wirkstoffe erfüllen müssen, ist die Erforschung sowohl des Wirkprinzips als auch der Pharmakokinetik des Wirkstoffes von besonderer Bedeutung. Obwohl Johanniskrautextrakte bereits seit dem 17. Jahrhundert gegen die Melancholie und somit als Antidepressivum eingesetzt wurden, sind sowohl der exakte Pathomechanismus der Depression als auch die Wirkkomponente und der Wirkmechanismus der Extrakte aus Hyperici herba noch immer Gegenstand umfassender Forschung. Vor allem wegen der geringen Nebenwirkungsrate sind Johanniskrautpräparate gegenüber synthetischen Antidepressiva eine bevorzugte Alternative für die Therapie leichter bis mittelschwerer Depressionen. Ähnlich den Effekten der synthetischen Antidepressiva sind Johanniskrautextrakte in der Lage, durch eine Wiederaufnahmehemmung der Neurotransmitter Noradrenalin, Serotonin, Dopamin, sowie GABA und L-Glutamat deren Konzentration im synaptischen Spalt zu erhöhen, was zu einer nachfolgenden adaptiven Veränderung der jeweiligen Rezeptoren führt. In mehreren Studien konnten diese Effekte vornehmlich den Phloroglucinolderivaten Hyperforin und Adhyperforin zugeordnet werden. Metabotropic glutamate receptor subtype 7 (mGluR7) belongs to the family of G-protein coupled receptors. mGluR7 is widely distributed in the brain and primarily localized at presynaptic terminals, where it is thought to regulate neurotransmitter release and synaptic plasticity. Studies have shown that the intracellular C-terminal tail of mGluR7 binds a variety of proteins in addition to trimeric G-proteins. These newly identified protein interactions are believed to play a key role in the synaptic targeting and G-protein dependent signaling of mGluR7. Protein interacting with C kinase 1 (PICK1), a PDZ-domain protein, is a strong interaction partner of mGluR7a. In order to investigate the role of PICK1 in the synaptic trafficking and signaling of mGluR7a, a knock-in mouse line in which the interaction of mGluR7a and PICK1 is disrupted was generated. Analysis of the mutant mice by immunocytochemistry and immunoelectron microscopy showed that the synaptic targeting and clustering of mGluR7a was not altered, indicating that PICK1 is not required for mGluR7a receptor membrane trafficking and synaptic localization. However, when the spontaneous synaptic activity of cerebellar granule cell cultures prepared from both wild-type and knock-in mice was monitored, and L-AP4 (400μm) was found to decrease the frequency, but not the amplitude, of spontaneous excitatory currents in wild-type neurons, while no effect of L-AP4 on spontaneous synaptic activity was observed in knock-in neurons. This indicates that PICK1 binding to the C-terminal region of mGluR7a plays an essential role in mGluR7a mediated G-protein signaling. We examined the threshold sensitivity for the convulsant pentetrazole (PTZ) in knock-in mice. It was found that mGluR7a knock-in mice had a greater sensitivity to PTZ than wild-type mice. Moreover, the surface parietal cortex EEG recordings of the mutant mice revealed spontaneous synchronous oscillation, or "spike-and-wave discharges" (SWD), which displayed similar characteristics to absence-like seizures. It was also observed that the knock-in mice responded to pharmacology as human absence epilepsy. These data suggests that the knock-in mice displayed the phenotype of absencelike epilepsy. Furthermore, the behavioral analysis of the mGluR7a knock-in mice showed no deficits in motor coordination, pain sensation, anxiety as well as spatial learning and memory, thus the interaction of mGluR7a and PICK1 appears not to contribute to these physiological processes. Taken together, our data provides evidence for an important role of PICK1 in Gprotein dependent signaling of mGluR7a, whereas PICK1 is not required for synaptic targeting and clustering of mGluR7a. Our results also provide an animal model of absencelike epilepsy generated by disruption of a single mGluR7a-PDZ interaction, thus creating a novel therapeutic target against this neurological disease.
Glutamat ist der häufigste Neurotransmitter im menschlichen Hirn. Die Konzentration des Glutamats in der extrazellulären Flüssigkeit wird durch Glutamat-Transporter (Sekundärtransporter) kontrolliert. Liegt es in zu hoher Konzentration im synaptischen Spalt vor, kommt es zur Schädigung von Nervenzellen, ein Prozess, der als Exzitotoxizität bezeichnet wird. Eine Fehlfunktion oder fehlerhafte Produktion der Glutamat-Transporter im zentralen Nervensystem wird bei verschiedenen Krankheiten, wie der amyotrophen Lateralsklerose, der Ischämie, der Epilepsie, der Schizophrenie und der Alzheimer-Krankheit vermutet. Ziel dieser Arbeit war die Funktions- und Strukturanalyse der Glutamat-Transporter GLT-1 aus Rattus norvegicus und GltP aus E. coli, um die Familie der Glutamat-Transporter und die Entstehung der mit diesen Transportern in Verbindung gebrachten Krankheiten besser zu verstehen. Um die für diese Analysen gebrauchten Mengen an Protein herzustellen, mussten die Proteine heterolog produziert werden, da sie in natürlichen Geweben nicht in ausreichender Menge vorkommen. In dieser Arbeit wurde Glutamat-Transporter GLT-1 aus Rattus norvegicus funktional mit dem Semliki Forest Virus Expressionssystem überproduziert. Dazu wurden verschiedene Vektorkonstrukte hergestellt. Die routinemäßige Überproduktion des Transporters wurde im 8 l - Maßstab durchgeführt. In Zellen, die für die Produktion von GLT-1 mit rekombinanten, aktiven SF-Viren infiziert wurden, konnte eine sehr hohe Aktivität des Glutamat-Transporters nachgewiesen werden. Die Menge des hergestellten GLT-1 wurde in Bindungsexperimenten mit (2S,4R)-4-Methylglutamat quantifiziert: jede Zelle enthielt 3,5 x 106 Transporter: 61,04 pmol GLT-1/mg Gesamtprotein. Das entspricht einer Ausbeute von etwa 2-3 mg/8 l Zellkultur. Die hier durchgeführte Überproduktion des GLT-1-Glutamat-Transporters ist die erste Überproduktion eines eukaryotischen Sekundärtransporters mit dem Semliki Forest Virus Expressionssystem, bei dem große Mengen an aktivem Protein hergestellt werden konnten. Zudem ist die Ausbeute an funktionalem GLT-1 mit 61 pmol/mg Gesamtprotein verglichen mit den in der Literatur vorliegenden Daten zur Überproduktion eukaryotischer sekundärer Transporter mit anderen Expressionssystemen die höchste, die bis dato erreicht werden konnte. Der größte Anteil des heterolog produzierten GLT-1 war glykosyliert. Die gelelektrophoretische Analyse des aufgereinigten Transporters ergab zwei Banden, die ein apparentes Molekulargewicht von etwa 70-75 kDa und etwa 53-58 kDa hatten. In einer Western-Blot-Analyse konnten beide Banden des GLT-1-Transporters mit einem anti-His-Antikörper und einem anti-GLT-1-Antikörper nachgewiesen werden. Durch Deglykosylierung mit PNGase F und einer Trennung beider Banden durch Lektin-Affinitätschromatographie konnte gezeigt werden, dass es sich bei der 70-75 kDa-Bande um die glykosylierte Form und bei der 53-58 kDa-Bande um die nicht glykosylierte Form des Glutamat-Transporters handelte. Es wurde gezeigt, dass zwischen der Aktivität des GLT-1 und dessen Glykosylierung kein Zusammenhang besteht. Denn beide Formen lagen als vollständige, funktionale Transporter vor und transportierten nach Rekonstitution in Liposomen Glutamat. Der prokaryotische Glutamat-Transporter GltP aus E. coli wurde in dem E. coli-Stamm C43 (DE3) überproduziert. Die Ausbeute war etwa 2 mg pro Liter Kultur. Die Funktionalität des Transporters nach Rekonstitution in Lipidvesikel wurde durch spezifische Aufnahme von Glutamat gezeigt. Für die Solubilisierung beider Transporter aus den Zellmembranen wurden verschiedene Detergentien getestet. GltP ließ sich am besten mit DM oder DDM aus der Membran extrahieren, für die Solubilisierung des GLT-1 wurde mit großer Effizienz DDM oder CYMAL-7 eingesetzt. GltP und GLT-1 wurden mit einer Ni2+-NTA-Affinitätschromatographie in großer Menge und hoher Reinheit angereichert werden. Die Aufreinigungsprozedur beeinträchtigte nicht die Funktionalität des prokaryotischen GltP. Bei dem eukaryotischen Transporter GLT-1 war nach der Ni2+-NTA-Säule keine Transportaktivität mehr messbar. Durch Zusatz von Asolectin in den Wasch- und Elutionspuffern während der Aufreinigung konnte die Funktionalität des Transporters jedoch erhalten werden. Aufreinigungen mit anderen Lipiden unter anderem in Kombination mit Cholesterin lieferten einen Glutamat-Transporter, der in seiner Konformation stabilisiert, jedoch nach Rekonstitution nicht aktiv war. Eine weitere Steigerung der Ausbeute an aktivem GLT-1 konnte durch den Einsatz von Reduktionsmitteln, wie DTT oder b-Mercaptoethanol, die die Aggregation des Transporters verhinderten, erreicht werden. GltP katalysiert den elektrogenen Transport von Glutamat bzw. Aspartat unter Symport von mindestens zwei Protonen. GLT-1 transportiert ein Molekül Glutamat zusammen mit drei Na+-Ionen und einem Proton im Austausch gegen ein K+-Ion. Durch Transportmessungen konnte der hochspezifische Glutamat-Transport der aufgereinigten Transporter belegt werden. Der Glutamat-Transport des in Liposomen rekonstituierten GltP zeigte eine klare Abhängigkeit von einem anliegenden Protonengradienten. Aufgereinigtes und rekonstituiertes GLT-1 transportierte nur Aspartat bzw. Glutamat, wenn ein Na+ und ein K+-Gradient vorhanden waren. Die Aspartat- bzw. Glutamat-Aufnahme konnte bei beiden Transportern durch den kompetitiven nichttransportablen Inhibitor (2S,4R)-4-Methylglutamat blockiert werden. Der Assoziationsgrad der Glutamat-Transporter GltP und GLT-1 und das Gleichwicht zwischen den verschiedenen oligomeren Zuständen wurde in dieser Arbeit eingehend mit biochemischen Methoden untersucht: 1. „Cross-linking“-Studien, 2. Blaue Nativgelelektrophorese, 3. Analytische Ultrazentrifugation, 4. Laserlichtstreuung, 5. Gelfiltrationschromatographie. Die dabei erhaltenen Ergebnisse bewiesen eine tetramere Assoziierung beider Proteine. Die Gelfiltrationsexperimente zeigten, dass die Transporter in Detergenzlösung in unterschiedlichen Assoziationsgraden vorliegen. Das Gleichgewicht zwischen den oligomeren Formen war reversibel und abhängig von der Art und Konzentration des Detergenz, der Proteinkonzentration und der Temperatur. Zur Untersuchung der Struktur der Glutamat-Transporter wurden vor allem mit GltP zahlreiche 2D-Kristallisationsexperimente durchgeführt. Trotz Variation aller denkbar möglichen Parameter konnten keine Kristalle erhalten werden. Das beste Ergebnis war ein guter Einbau des Proteins in Lipidvesikel (etwa 80%). Da keine Kristalle erhalten wurden, wurde für beide Proteine eine Einzelpartikelanalyse durchgeführt. Dabei wurde nach zweidimensionaler Alignierung und Klassifizierung die „random conical tilt“-Methode angewendet. Die daraus resultierenden dreidimensionalen Dichtekarten des GltP und GLT-1 waren sehr ähnlich und wiesen vier nicht exakt symmetrische Massen in annähernd quadratischer Anordnung auf. Die Auflösung war 26 Å bzw. 36 Å. Die Größe der Einzelpartikel (für GltP: Höhe 37 Å, Breite 75 Å bzw. 86 Å, Länge 100 Å). ihre annähernd quadratische Anordnung und ihre Symmetrie lassen vermuten, dass es sich dabei um Tetramere der Glutamat-Transporter handelt, die aus zwei nicht symmetrischen Dimeren zusammengesetzt sind. Die hier präsentierten Daten sind die ersten zur dreidimensionalen Struktur von Glutamat-Transportern. Schließlich wurde nachgewiesen, dass der in BHK-Zellen heterolog exprimierte Glutamat-Transporter GLT-1 vorwiegend in „lipid rafts“ lokalisiert ist. Die Größe der „rafts“, die anhand der Größe der „Proteininseln“ in Gefrierbrüchen bestimmt wurde, war etwa 200 nm im Durchmesser. Die „GLT-1-Inseln“ bzw. „lipid rafts“ konnten durch das teilweise Entfernen von Cholesterin aus der Membran zerstört werden. Damit ging eine Reduktion der Glutamat-Transporter-Aktivität von etwa 20% einher. Es ist das erste Mal, dass „lipid rafts“ durch die natürliche Assemblierung von Proteinen mit Hilfe von Gefrierbruchanalysen und Elektronenmikroskopie beobachtet wurden.
Safety concerns associated with the use of viral vectors in gene therapy applications have attracted considerable attention towards the development of nonviral vectors as alternatives for DNA delivery. While nonviral vectors are commonly not associated with safety problems, they are still very inefficient compared to viral vectors, and require significant improvements to approach the efficiency of their viral counterparts. Meanwhile ligands or single-chain antibody fragments that bind to cell surface receptors for increased and/or specific cellular uptake, endosome escape activities, and nuclear localization sequences (NLSs) to enhance transport of plasmid DNA into the nucleus, have become available that can be incorporated into nonviral vectors to improve their efficacy. However, as gene delivery is a multistep process, the challenge is to incorporate multiple of these functional elements into a single nonviral vector system, while retaining their specific activities. A promising method to attach such entities to plasmid DNA is the use of multifunctional fusion proteins that bind to DNA through a DNA-binding domain. In principle, two types of DNA-binding domains/proteins can be used to anchor additional functional domains or peptides to a plasmid, namely sequence-specific DNA-binding domains, described in the first part of this thesis, or those that bind DNA independent of its sequence, exemplified in the second part of this work by a derivative of the human HMGB2 protein. The first fusion protein constructed and analyzed contained the E. coli LexA repressor as a sequence-specific DNA-binding domain. In addition, this DNA-carrier protein, termed TEL, included a bacterial translocation domain as an integrated endosome escape activity, and human TGF-a for specific targeting to the EGF-receptor (EGFR). TEL was expressed in E. coli and purified under both native and denaturing conditions. Purified, denatured TEL was refolded and subsequently shown to bind specifically to EGFR-expressing cells. However, inclusion of TEL in complexes of plasmid DNA and poly-L-lysine (pL) did not lead to increased gene delivery into EGFR-expressing COS-1 cells. Most likely this was due to the absence of DNA-binding activity of the LexA moiety in TEL. In contrast, native TEL was able to interact specifically with DNA. Nevertheless, since this interaction was rather weak, and refolding of denatured TEL had not resulted in functional activity of all of its protein domains, it seemed unlikely that fusion proteins containing LexA would exhibit gene transfer capabilities superior to those of similar DNA-carrier proteins previously constructed in our group. Further work therefore focused on the use of the E2C-Sp1C protein as an alternative sequencespecific DNA-binding domain. This artificial zinc-finger protein was fused to the single-chain antibody fragment scFv(FRP5), directed against the human ErbB2 growth factor receptor. The resulting 5-E2C fusion protein was expressed in E. coli and purified under native and denaturing conditions. Refolded and native 5-E2C were found to bind specifically to ErbB2-expressing cells, indicating that scFv(FRP5) in 5-E2C was functional in both preparations. In contrast, whereas refolded 5-E2C bound DNA only weakly, significant DNA binding was observed for native 5-E2C. In addition, it could not only be shown that the interaction of native 5-E2C with DNA containing its recognition sequence was specific, but also that this protein was able to bind DNA and recombinant ErbB2 simultaneously, demonstrating the functionality of both domains in native 5-E2C. Despite these encouraging results, the inclusion of native 5-E2C in pL- or polyethyleneimine (PEI)-DNA complexes did not lead to an (5-E2C-specific) enhancement of gene transfer efficiency, irrespective of the presence of the endosome-disruptive reagent chloroquine during transfection. In the second part of this thesis an alternative approach for the development of DNA-carrier proteins for nonviral gene delivery is described, based on human HMGB2, a DNA-binding protein without sequence specificity. HMGB2 contains an acidic C-terminus that has been found to decrease the affinity of the protein for DNA. Therefore, this C-terminal tail was deleted, resulting in an HMGB2-variant consisting of amino acids 1-186. HMGB2186, purified under native conditions from E. coli lysates, was able to interact with DNA and bound to the surface of different cell lines. Importantly, after binding to plasmid DNA HMGB2186 mediated gene delivery into COS-7 cells with higher efficiency than pL. In addition, HMGB2186-mediated gene transfer was strongly enhanced in the presence of chloroquine, indicating that the endocytic pathway was involved in cellular uptake. To improve internalization and intracellular routing of HMGB2186 as a DNA-carrier, a derivative containing the TAT47-57 cell-penetrating peptide (CPP), reported to facilitate cell entry independent of endocytosis, was constructed. Since this peptide also contains an NLS, in addition an HGMB2186-variant containing the SV40-NLS was constructed to investigate the effect of a peptide that has only nuclear localizing properties. Interestingly, the resulting TAT-HMGB2186 and SV40-HMGB2186 fusion proteins displayed DNA-binding activities similar to HMGB2186, but mediated gene delivery into different cell lines clearly more efficiently than the parental molecule. Furthermore, the efficacy of both fusion proteins was enhanced markedly in the presence of chloroquine, an indication that endocytosis was involved in the transfection process mediated by these proteins. This suggests that the increased transfection efficiency observed for TAT-HMGB2186 was more likely due to the NLS function present in the TAT47-57 peptide, rather than to its ‘cell penetrating properties’. Finally, the incorporation of functional peptides derived from human proteins into HMGB2186 was investigated. An uncharged CPP originating from Kaposi-FGF, reported to facilitate efficient cellular uptake of fused protein domains in an endocytosis-independent manner, was fused to HMGB2186 together with the SV40-NLS. Interestingly, the resulting KSV40-HMGB2186 fusion protein bound DNA similarly as previously tested DNA-carrier proteins, but did not mediate enhanced transfection compared to HMGB2186. In addition, the importin-b-binding (IBB) domain derived from human importin-a2 was investigated as a component of a DNA-carrier protein. Since the IBB domain can function as an NLS, it was fused to HMGB2186 resulting in the DNA-carrier protein IBBHMGB2186. Although IBB-HMGB2186 bound DNA in a similar manner as the other HMGB2186-derivatives, gene delivery mediated by IBB-HMGB2186 was only as effective as HMGB2186 mediated transfection, suggesting no significant role of the IBB domain. However, addition of chloroquine resulted in a remarkable enhancement of IBB-HMGB2186-mediated gene transfer, which was now more efficient than with any other HMGB2186-variant tested, and not much lower than gene transfer mediated by PEI, one of the most efficient transfection reagents available to date. To enhance nonviral gene delivery even further, the HMGB2186-based DNA-carrier proteins described in this thesis might now serve as building blocks for novel fusion proteins that include additional complementing activities. In this respect it seems particularly promising that, under conditions of effective end some escape, IBB-HMGB2186, which consists entirely of protein domains of human origin, was the most efficient of all proteins tested in this work.
RcsB is a central transcriptional regulator in enteric bacteria involved in exopolysaccharide (EPS) biosynthesis, in cell division, in the expression of osmoregulated genes, and regulates at least 20 other genes and operons. It is a member of a phosphorelay system and signal transfer is mediated by phosphorylation through the RcsC/YojN phosphorelay. RcsB proteins modified with the phosphorylation mimic BeF3- as shown by its conformational changes and DNA binding properties and resulted phosphorylated RcsB derivatives with sufficient stability. Both, the wild type RcsB protein and the mutant RcsBD11A could be modified with BeF3-. Non-phosphorylated RcsB has been shown to bind as a heterodimer with the coinducer RcsA at the conserved RcsAB box in Rcs regulated promoters. In this study, it has been shown that the modification of RcsB by BeF3 - (I) has a negative effect on its homodimerization, (II) abolishes the complex formation of RcsAB with the RcsAB box as shown by the EMSA and SPR technique. All the effects were found to be reversible by increasing the NaF concentration in the assays presumably leading to the formation of the inactive BeF4 2- salt. This hypothesis of RcsB being modified by BeF3- was also supported by other phosphodonors like ATP and acetyl phosphate, both of them showed the same negative effect on DNA binding by RcsAB heterodimer giving evidence that BeF3- could be used as a phosphorylation mimic. In addition, the phosphorylation mimic BeF3- was found to be a better phosphorylating agent than ATP and acetyl phosphate. This is the first evidence that phosphorylation of RcsB might have a negative effect on the activation of RcsAB regulated operons. Autophosphorylation of RcsB proves that it has the ability to take up phosphoryl groups and the mutant protein also become autophosphorylated with less efficiency or stability than the wild type protein. RcsB probably takes up phosphoryl groups through RcsC -> YojN -> RcsB phosphorelay pathway. To study the interaction among the proteins in this pathway, fluorescence spectroscopy, NMR spectroscopy, and an in vivo ß galactosidase assay were performed by using two domains of RcsC (T-RcsC and R-RcsC), HPt domain of the protein YojN, and RcsB. The interactions between R-RcsC/YojN-HPt and YojN-HPt/RcsB supports the proposed pathway of phosphorylating RcsB. RcsB might also be phosphorylated by YojN-HPt that is phosphorylated by other sensor kinase other than RcsC in a cross-talk mechanism. The phosphorylation of RcsB by YojN-HPt probably has the same negative effect on cps induction as obtained with BeF3 - effect on DNA binding by RcsAB heterodimer.
Untersuchungen zur ZNS-Bioverfügbarkeit wirksamkeitsbestimmender Inhaltsstoffe von Johanniskraut

In der vorliegenden Arbeit wurde die ZNS-Bioverfügbarkeit der Quercetin-Flavone und der Naphthodianthrone aus Hypericum perforatum untersucht. Hierzu wurde jew. eine HPLC-gestütze Methode zur Quantifizierung der genannten Stoffe in ZNS und Plasma erstellt und in enger Anlehnung an internationale Richtlinien (ICH- u. FDAGuidelines) validiert. Mit Hilfe dieser Methoden wurden im Anschluss Rattenfütterungsstudien durchgeführt und ausgewertet. Im Falle der Quercetin-Flavone konnte eindeutig gezeigt werden, dass diese Stoffgruppe ZNS-bioverfügbar ist und bei wiederholter Gabe (1 x tgl.) über den beobachteten Zeitraum von 8 Tagen im ZNS kumuliert. Ferner ist es gelungen, erste ZNS-Spiegelkurven der Quercetin-Flavone nach Einmal- und Mehrfach-Gabe aufzuzeichnen. Im Gegensatz hierzu weisen die Naphthodianthrone, unter Berücksichtigung der analytischen Grenzen, keine ZNS-Bioverfügbarkeit auf. Betrachtet man diese Ergebnisse im Kontext der bisher erschienenen Publikationen zu Johanniskraut, lässt sich sagen, dass durch die vorliegende Arbeit die Rolle der Flavonoide eindeutig an Bedeutung gewinnt und die der Naphthodianthrone eindeutig an Bedeutung verliert. Schaut man auf die bisher untersuchten Inhaltsstoffe von Johanniskraut, wird deutlich, dass den Phloroglucinolen die wohl herausragendste Bedeutung zukommt, was nicht zuletzt durch Keller et al.[137] und Müller et al. [27,59,67] gezeigt werden konnte. Für die klinische Wirksamkeit des Hypericum-Extraktes sind jedoch auch die Flavonoide essentiell, was durch Tierstudien gezeigt und durch diese Arbeit unterstüzt werden konnte[114]. Der Mechanismus, durch den die Flavonoide zur antidepressiven Wirksamkeit beitragen, liegt, im Gegensatz zu dem der Phloroglucinole, jedoch noch im Dunklen. Durch die Präsenz von Quercetin bzw. dessen Metaboliten im ZNS auf der einen und den Ergebnissen von Tierstudien zur antidepressiven Wirksamkeit isolierter Flavonoide mit Hilfe des Porsolt-Tests auf der anderen Seite[33] lässt sich jedoch spekulieren, dass die Flavonoide einen direkten antidepressiven Effekt ausüben und nicht nur indirekt durch Verbesserung, beispielsweise der ZNS-Bioverfügbarkeit der Phloroglucinole o. ä., zu den antidepressiven Eigenschaften des Extraktes beitragen. Durch ihr Unvermögen, die Blut-Hirn-Schranke in nennenswerten Konzentrationen zu überschreiten, treten die Naphthodianthrone, die in der Vergangenheit als für die Wirksamkeit wichtigsten Inhaltsstoffe angesehen wurden, eindeutig in den Hintergrund. Als Beitrag dieser Stoffgruppe zur antidepressiven Wirksamkeit ist demnach erstens ein wie auch immer gearteter indirekter Effekt durch Verbesserung der ZNS-Bioverfügbarkeit der ZNS-gängigen Substanzen, zweitens ein wegen der bestenfalls möglichen, niedrigen Konzentrationen schwacher antidepressiver Effekt im ZNS, drittens ein antidepressiver Effekt durch aktive Metaboliten oder viertens ein antidepressiver Effekt, der sich in der Peripherie beispielsweise durch Beeinflussung der HPA-Achse abspielt, denkbar. Für ersteres existieren bis dato keinerlei Anhaltspunkte, für den zweiten Punkt existieren weder Beweis noch Gegenbeweis noch Spekulationen in der Literatur, dasselbe gilt für den dritten Punkt, der vierte Punkt wird hingegen kontrovers diskutiert[44,97,143]. In Publikationen wurde hierzu festgestellt, dass unter dem Einfluss von Hypericin sich die Blutspiegel an Cortisol und ACTH, die bei Depressiven erhöht bzw. gestört sind, wieder normalisieren. Der Einwand der Kritiker dieses Wirkmodells scheint hierbei aber plausibel, da es sich bei dem mit Hilfe von Hypericin beeinflussbaren, beobachteten Phänomen der Erhöhten Cortisol- und ACTH-Plasmaspiegel auch nur um ein Symptom einer Depression und nicht um einen für die Depression ursächlichen Vorgang handeln kann. Nicht jeder, bei dem erhöhte Cortisol- und veränderte ACTH-Spiegel vorliegen ist zwangsläufig depressiv. Das exakte Zusammenspiel der einzelnen Extrakt-Komponenten, von denen die bisher in den Focus der Untersuchungen geratenen Stoffe nur etwa 10 bis 15% ausmachen, bleibt jedoch auch weiterhin zum größten Teil im Unklaren, wenngleich gerade dieses Zusammenspiel die besonderen Eigenschaften des Extraktes und dessen Wirkung ausmachen. Unverzichtbare Bestandteile sind hierbei Phloroglucinole und Flavonoide, die in angemessenen Konzentrationen im Extrakt repräsentiert sein müssen, um ein klinisch hochwertiges Produkt zu gewährleisten. Inwieweit ein repräsentativer Gehalt an Hypericinen im Extrakt vorhanden sein sollte, ist nicht zuletzt auf Grund der Ergebnisse dieser Arbeit mit einem Fragezeichen zu versehen, wenngleich die Datenlage zur Zeit keinesfalls ausreicht, die Hypericine gänzlich aus dem Extrakt zu verbannen. In wie weit die An- oder Abreicherung einzelner Komponenten zu einer Verbesserung der klinischen Wirksamkeit führt, muss jedoch stets in zusätzlichen Studien eruiert werden. Insgesamt steht aber mit dem Johanniskraut-Extrakt für die erfolgreiche Therapie von milden bis mittelschweren Depressionen ein wichtiges, fast unverzichtbares Werkzeug zur Verfügung, das sich insbesondere durch sein verglichen mit synthetischen Antidepressiva, überaus günstiges Profil an unerwünschten Arzneimittel-Wirkungen auszeichnet. Trotz in jüngster Zeit propagierter Bedenken hinsichtlich der Unbedenklichkeit der Anwendung von Johanniskraut Extraktpräparaten besonders in Kombination mit anderen Arzneistoffen, was jedoch einer genaueren Betrachtung nur schwer standhält[144], stellt der Johanniskraut-Extrakt zur Behandlung milder bis mittelschwerer Depressionen ein unverzichtbares Werkzeug dar.
P2X receptors are ligand (ATP)-gated ion channels that open an intrinsic cation permeable pathway in response to extracellular ATP released from both neuronal and non-neuronal cells. P2X receptors are abundantly distributed and mediate a wide variety of physiological functions, ranging from fast synaptic transmission in the central, peripheral, and enteric nervous system, to proinflammatory cytokine release from immune cells. The primary aim of this work was to elucidate the pathway that leads to the finally assembled trimeric P2X receptors, including the assessment of a possible role of ER chaperones and folding factors in this process. Additionally, the study was conducted to investigate the various ER quality control processes involved in the selection of “properly folded and assembled” P2X receptors that are suitable for the surface expression.
Tumorspezifische T-Zellen stellen den im Prinzip wirkungsvollsten Mechanismus zur Abstoßung von Tumoren dar. Voraussetzung für die Aktivierung tumorspezifischer zytotoxischer CD8+ T-Zellen (Tc oder CTLs) ist die Präsentation von Tumorantigenen auf MHC-Klasse I Molekülen durch professionelle Antigen-präsentierende Zellen (APCs). Wegen ihrer Schlüsselrolle bei der Aktivierung von T-Zellen werden APCs in der aktiven Krebs-Immuntherapie eingesetzt. Unabhängig von CTLs können auch CD4+ Helfer T-Zellen (Th) eine Reihe effektiver Anti-Tumor Mechanismen einleiten. Die Aktivierung von CD4+ erfordert die Präsentation des Antigens via MHC-Klasse II Molekülen. Bis jetzt konnten jedoch nur einige wenige tumor-spezifische Th-Epitope identifiziert werden, welche auf MHC-Klasse II Molekülen präsentiert werden. Aus diesem Grund benutzen die meisten derzeitigen Vakzinierungsstrategien definierte Tc-Epitope, die in der Lage sind, spezifische Immunantworten gegen das Tumorantigen auszulösen. Diese Vakzine basieren auf tumorspezifischen Peptiden, Proteinen oder Proteinfragmenten, aber auch auf DNA-Konstrukten, welche für ein bestimmtes Tumorantigen kodieren. Um allerdings eine ausreichend starke Immunreaktion auszulösen, ist es notwendig diese normalerweise nur schwach immunogenen Substanzen zusammen mit Adjuvanzien zu verabreichen. Dendritische Zellen (DCs) sind hoch-effiziente APCs. Weil sie in der Lage sind, die primäre Immunantwort einzuleiten werden sie auch als „natürliches Adjuvanz“ bezeichnet. Nicht zuletzt deswegen werden bei der Entwicklung von Vakzinen immer mehr die einzigartigen Eigenschaften von DCs ausgenutzt. Gegenwärtige Studien untersuchen daher die Beladung von DCs mit synthetischen, tumorspezifischen Peptiden oder Tumor-Lysaten, um eine Aufnahme des Tumorantigens sowie dessen anschließende Präsentation auf MHCKlasse I Molekülen zu erreichen. Obgleich einige dieser Vakzinierungs-Ansätze zu vielversprechenden Ergebnissen führten, sind solche Strategien mit der aufwendigen und kostenintensiven ex vivo Aufbereitung primärer Dendritischer Zellen verbunden. Zudem muss gewährleistet sein, dass die ausgewählten T-Zell Epitope auch vom jeweiligen MHC-Haplotyp des Patienten präsentiert werden. Um einige dieser Nachteile auszugleichen, wären „Konstrukte“ wünschenswert, welche in vivo einen zielgerichteten Transport eines oder besser, mehrerer T-Zell Epitope zu den Dendritischen Zellen ermöglicht. Mit der Entwicklung und Optimierung von Systemen, die Tumorantigene gezielt zu und in APCs transportieren, könnte die Effizienz herkömmlicher Vakzinierungen gesteigert und deren Dosierung verringert werden. In dieser Arbeit wird die Herstellung sowie die funktionelle Charakterisierung zweier neuartiger Vakzine beschrieben. Diese Vakzine wurden für die in vivo Vakzinierung gegen das tumorassoziierte Antigen ErbB2 (HER2/neu) entwickelt. ErbB2 wird in einer Reihe von Tumoren wie Brust- und Ovarialkarzinomen oder Adenokarzinomen der Lunge überexprimiert. Andere Tumorenentitäten wie Kolon-, Nieren-, Blasen-, Speicheldrüsen-, Prostata- und Pankreaskarzinome zeigen ebenfalls eine variable Überexpression von ErbB2. Ziel des ersten der beiden Vakzinierungsansätze war eine verbesserte Aufnahme und Präsentation von Tumorantigenen durch APCs zu erreichen. Als Teil eines Fusionsproteins sollte hier die zielgerichtete Bindung einer antigenen Determinante an APC-spezifische Oberflächenmoleküle und deren anschließende Aufnahme und Prozessierung innerhalb von APCs ermöglicht werden. Als mögliche APC-Bindedomäne wurde ein Fragment gewählt, welches sich vom extrazellulären Anteil des B7-Rezeptors CTLA-4 (CTLA-4125) ableitet. B7 ist ein Oberflächenprotein, das speziell auf APCs exprimiert wird. Nach der bakteriellen Expression und Reinigung wurde durch FACS-Analyse gezeigt, dass lösliches humanes CTLA-4125 spezifisch sowohl an murine als auch humane B7 Moleküle bindet und daher als effektive APC-Bindungsdomäne für den zielgerichteten Transport von Tumorantigenen zu APCs verwendet werden kann. Als chimäre Proteinvakzine zur spezifischen Bindung an APCs wurden zwei mit C-ErbB2(N) bzw. C-E-ErbB2(N) bezeichnete Fusionsproteine konstruiert. Diese setzen sich aus CTLA-4125 am N-Terminus für die Bindung an APCs und einem Fragment des extrazellulären Teils des humanen tumorassoziierten ErbB2 Antigens (ErbB2(N), Aminosäuren 1-222) am C-Terminus zusammen. Beim C-E-ErbB2(N) Protein wurde zusätzlich die Translokationsdomäne von Pseudomonas exotoxin A (ETA II) zwischen der APC-Bindungsdomäne und der antigenen Determinante eingefügt. Diese Domäne könnte nach Rezeptor-vermittelter Aufnahme des Fusionsproteins die Freisetzung des Tumorantigens aus dem Endosom ins Zytoplasma verbessern und so die Prozessierung und Präsentation des Antigens via MHC-Klasse I verstärken. Beide chimären CTLA-4-ErbB2 Fusionsproteine wurden in E. coli exprimiert und mittels Metallchelat-Chromatographie aus den bakteriellen Lysaten gereinigt. Die spezifische Bindung der rekombinanten Fusionsproteine an B7 Moleküle auf der Zelloberfläche sowie deren Aufnahme durch B7-exprimierende humane Raji Zellen wurde mittels FACS Analyse und konfokaler Mikroskopie nachgewiesen. In Balb/c Mäusen wurde untersucht, inwieweit die Fusionsproteine eine Immunantwort gegen das Tumorantigen auslösen können. Die Vakzinierung von Balb/c Mäusen mit den CTLA-4 Fusionsproteinen bewirkte die Abstoßung anschließend subkutan injizierter syngener ErbB2-positiver Nierenkarzinomzellen (Renca-lacZ/ErbB2) nach subkutaner Injektion. Dies deutet auf die Erzeugung protektiver Immunität hin. Obgleich beide Fusionsproteine effektiv waren, zeigte das Fusionsprotein C-ErbB2(N), dem die bakterielle Translokationsdomäne fehlt, in mehreren unabhängigen Experimenten eine höhere Wirksamkeit als C-E-ErbB2(N). Vakzinierung mit den CTLA-4 Fusionsproteinen erzeugte keinen Schutz gegen ErbB2-negative Kontroll-Tumorzellen. Dies Experimente zeigt die Antigen-Spezifität der induzierten Immunantwort nach Vakzinierung mit CTLA-4 Fusionsproteinen. T-Zell Depletionsexperimente ergaben, dass die protektiven Effekte eindeutig von CD8+ CTLs abhängig waren. Eine erneute, diesmal intravenöse Applikation von ErbB2+ Tumorzellen in vakzinierte, tumorfreie Mäuse zwei Monate nach der ersten, subkutanen Injektion („Re-challenge“) führte nicht zur Ausbildung von Lungenmetastasen. Dies deutet darauf hin, dass durch die Vakzinierung eine langanhaltende Immunreaktion induziert wurde. In therapeutischen Vakzinierungsexperimenten, die wohl am ehesten eine klinische Situation in Tumorpatienten widerspiegeln, wurden die CTLA-4 Fusionsproteine in die Umgebung bereits etablierter subkutaner Renca-lacZ/ErbB2 Tumoren injiziert. In zwei unabhängigen Experimenten konnte gezeigt werden, dass durch die therapeutische Vakzinierung mit CErbB2(N) 7 von 8 Mäusen geheilt werden konnten, wohingegen die Wirkung von C-EErbB2(N) – von 8 Mäusen wurden 6 geheilt – auch im therapeutischen Einsatz etwas schwächer zu sein schien. Nach einem intravenösen „Re-challenge“ mit Renca-lacZ/ErbB2 Zellen waren bereits geheilte Tiere auch vor der Ausbildung von Lungenmetastasen geschützt. Dies bestätigt eine durch die Vakzinierung hervorgerufene lang-anhaltende Anti-Tumor Immunität. Alternativ zur Produktion rekombinanter Proteinvakzine in Bakterien wurde auch die direkte Injektion von DNA-Konstrukten zur in vivo Expression und Sekretion der CTLA-4 Fusionsproteine getestet (DNA-Vakzinierung). Basierend auf dem Plasmid pSecTag2 wurden Säugerzell-Expressionsvektoren hergestellt, welche unter der Kontrolle eines CMVPromotors die beiden Fusionsproteine C-ErbB2(N) bzw. C-E-ErbB2(N) kodieren. Ein Igk “leader” Signalpeptid ermöglicht hierbei die Sekretion der produzierten Proteine. Nach Transfektion der Plasmide in COS-7 Zellen konnte Sekretion der CTLA-4-Fusionsproteine in den Zellkulturüberstand sowie deren Zellbindungsspezifität für B7, mit Hilfe von Western-Blot und FACS-Analysen nachgewiesen werden. Protektionsexperimente ergaben, dass alle Balb/c Mäuse, die vorher durch intramuskuläre Injektion der CTLA-4-ErbB2 DNA-Vakzine vakziniert wurden, vor dem Anwachsen subkutan injizierter Renca-lacZ/ErbB2 Tumorzellen geschützt waren. Mäuse, die als Kontrolle mit dem leeren pSecTag2 Vektor vakziniert wurden, entwickelten dagegen schnell wachsende Tumoren. In diesem ersten Teil dieser Arbeit konnte gezeigt werden, dass chimäre Proteinvakzine, die in einer einzigen Polypeptidkette eine Zellerkennungsdomäne zur spezifischen Bindung an APCs und ein antigenes Proteinfragment aus dem humanen ErbB2 enthalten, in vivo eine äußerst wirkungsvolle ErbB2-spezifische T-Zell vermittelte Anti-Tumor Immunantwort auslösen. Hierbei konnte sowohl durch die Injektion von rekombinanten, in Bakterien hergestellten Proteinen, wie auch durch DNA-Vakzinierung zur in vivo Produktion dieser chimären Proteinvakzine erreicht werden, dass syngene, ErbB2 exprimierende Tumoren abgestoßen wurden. Die oben beschriebenen chimären Proteinvakzine enthalten ein großes Proteinfragment, abgeleitet aus einem humanen Tumorantigen, welches zu APCs befördert und von ihnen aufgenommen wird. Dieser Ansatz ist nicht unbedingt abhängig von einem bestimmten MHCTyp und möglicherweise besonders dann wirkungsvoll, wenn Immunantworten gegen Antigene erzeugt werden sollen, für die noch keine von MHC-Molekülen präsentierten Peptide identifiziert sind. Für Tumorantigene, aus welchen bereits einzelne Peptide als potentielle Tumorabstoßungs-Antigene definiert sind, wurden in einzelnen Fällen synthetische Derivate solcher Peptide erfolgreich eingesetzt, um hochspezifische T-Zell Antworten zu erzeugen. Aber auch in diesem Fall könnte die Verknüpfung des Antigens mit einem geeigneten „Antigen-Carrier“ eine verbesserte Aufnahme und Präsentation durch APCs und eine Verstärkung der Immunantwort gegen das Antigen zur Folge haben. Aus diesem Grund wurde in einem alternativen Ansatz zur Induktion einer ErbB2-spezifische Anti-Tumor Immunität die in vivo Anwendung von synthetischen Vakzin-Komplexen untersucht, welchen ein liposomaler Träger als Vektor für Peptid-Antigene zugrunde liegt. Diese Peptide sind über einen immunstimulatorischen Lipopeptid-Anker (Pam3CysSerSer) mit der Oberfläche solcher Liposomen verknüpft. Dieser Teil der Arbeit wurde in enger Kollaboration mit Audrey Roth, Dr. Benoit Frisch und Dr. Francis Schuber, Laboratoire de Chimie Bioorganique, Faculté de Pharmacie, Université Louis Pasteur (ULP), Strasbourg ausgearbeitet, die alle in dieser Arbeit verwendeten liposomalen Vakzine konstruierten und für in vivo Untersuchungen bereitstellten. Zwei liposomale Substanzen wurden synthetisiert. Ein Mono-Epitop Konstrukt, als „Tc-ErbB2-liposomes“ bezeichnet, trägt ein Tc-Epitop (TYLPTNASL), welches sich von der Aminosäuresequenz des humanen ErbB2 Moleküls ableitet. Es war bereits vorher gezeigt worden, dass dieses Peptid mit hoher Affinität an das murine H-2Kd MHC-Klasse I Molekül bindet und eine Peptid-spezifische CTL Immunantwort in Balb/c Mäusen auslösen kann. Die zweite liposomale Substanz, als „Tc-ErbB2/Th-liposomes“ bezeichnet, enthält zusätzlich zum ErbB2-Peptid noch ein universelles Th-Epitop, das sich vom Influenza Virus Hämagglutinin (HA 307-319) ableitet und ebenfalls über den adjuvanten Anker mit dem Liposom verbunden ist. Durch das Hinzufügen des Th-Epitops wurde beabsichtigt, zusätzlich CD4+ Helfer TZellen zu rekrutieren und damit die CTL Antwort gegen das Tc-Epitop zu verstärken. Durch ex vivo Restimulation von Milzzellen aus zuvor mit den liposomalen Konstrukten vakzinierten Tieren mit dem ErbB2-Peptid konnte die in vivo Aktivierung Tc-spezifischer CTLs nachgewiesen werden. In dem gleichen immunkompetenten Balb/c Maus-Modell, das auch zur Charakterisierung der CTLA-4-ErbB2 Proteinvakzine verwendet worden war, wurde auch die anti-tumorale Aktivität der liposomalen Komplexe untersucht. Die Vakzinierung von Balb/c Mäusen mit beiden Konstrukten, Tc-ErbB2-liposomes und Tc-ErbB2/Th-liposomes, führte zur Abstoßung anschließend subkutan injizierter ErbB2 exprimierender RencalacZ/ErbB2 Zellen. Eine therapeutische Vakzinierung von Mäusen, die bereits etablierte ErbB2+ Tumoren trugen, führte zu einem verlangsamten Wachstum in einigen und zur kompletten Abstoßung in den übrigen Tieren. In Kontrolltieren, die lediglich mit dem liposomalen Träger vakziniert waren, konnten keinerlei Tumorregression beobachtet werden. Während beide liposomalen Substanzen sowohl bei der protektiven als auch bei der therapeutischen Vakzinierung Anti-Tumor Effekte zeigten, verstärkte das Hinzufügen des Th-Epitops zu den „Tc-ErbB2 liposomes“ sowohl die ErbB2-spezifische Immunantwort als auch die Anti-Tumor Immunität. Diese Ergebnisse unterstreichen die Bedeutung einer synergistischen CD4+ T-Zell Aktivierung als Teil von Anti-Tumor Immunantworten. Unabhängig von ihrer unterschiedlichen Wirkungsweise und chemischen Zusammensetzung stellen beide in dieser Arbeit untersuchten Vakzinkomplexe effektive Tumorvakzine dar, die nach ihrer Anwendung in vivo ErbB2-spezifische, T-Zell vermittelte Anti-Tumor Immunantworten auslösen können. Die chimären Fusionsprotein-Vakzine enthalten ein großes antigenes Proteinfragment. Durch ihre spezifische Zellbindungsdomäne wird der zielgerichtete Transport zu und die Aufnahme des Antigens in APCs, gefolgt von in vivo Prozessierung des Proteins in kleine antigene Peptide, MHC-Präsentation und schließlich Aktivierung tumorspezifischer T-Zellen gewährleist. Dieser Ansatz könnte insbesondere dann nützlich sein, wenn für das jeweilige Tumorantigen noch keine passenden T-Zell Epitope charakterisiert sind. Ein zusätzlicher Vorteil dieser Konstrukte ist, dass in einem einzigen Vakzin mehrere unterschiedliche antigene Determinanten bereitgestellt werden können. Für eine Reihe von Tumorantigenen sind bereits einzelne Peptide als potentielle Tumorabstoßungs-Antigene charakterisiert worden. Die in dieser Arbeit beschriebenen neuartigen liposomalen Träger, welche zusätzlich zum Tc-Epitop einen immunstimulatorischen Anker und ein starkes Th-Epitop tragen, könnten dazu beitragen, die Immunogenität und die anti-tumorale Aktivität synthetischer Peptid-Vakzine zu erhöhen.
Das Genom des anaeroben ε-Proteobakteriums Wolinella succinogenes codiert überraschenderweise für Enzymkomplexe, die typisch für die aerobe Atmung sind. Die entsprechenden Gene werd en unter bekannten Wachstumsbedingungen nicht
exprimiert. Darunter findet sich ein Operon ( sdhABE) für eine putative „Succinat-Dehydrogenase“, die hohe Sequenzhomol ogien zu sog. „nicht-klassischen“ archaealen Succinat-Dehydrogenasen zeigt. In der vorliegenden Arbeit sollte die „Succinat-Dehydro genase“ mit Hilfe des etablierten genetischen Systems zur Modifikation und Produktion der Chinol:Fumarat-Reduktase(QFR) homolog in W. succinogenes produziert und charakterisiert werden. Das genetische System besteht aus der QFR- Deletions-mutante ΔfrdCAB und dem Vektor pFrdcat2, der aufgrund seiner zentralen Bedeutung im Rahmen dieser Arbeit sequenziert wurde.
Summary and Outlook The aim of this work was the investigation of the Mn2+ binding sites in hammerhead and the Diels-Alder ribozymes. This project consists of three main topics. In the first part quantification and structural characterization of Mn2+ binding sites in the m- and the tsHHRz using Electron Paramagnetic Resonance (EPR) spectroscopy are described. The second part summarizes the newest results obtained for the cleavage activity of both mand tsHHRzs in the presence of different Mg2+ and Mn2+ and Na+ ion concentrations using the new method with fluorescent-labeled RNAs. Here the influence of neomycin B on the structure of Mn2+ binding pockets and on the catalytic activity of both HHRzs is discussed. In addition, a possible role of Mn2+ ions is suggested from correlation of the EPR data with the kinetic results. The last chapter is devoted to quantification and differentiation of Mn2+ binding sites of the Diels-Alder ribozyme using continuous wave (cw) EPR experiments in solution. In this work EPR spectroscopy was used to study the binding of Mn2+ ions to the cis tsHHRz and to compare it with the binding to the trans mHHRz and to the Diels-Alder ribozyme. Cw EPR measurements showed that the tsHHRz possesses a single highaffinity Mn2+ binding site with a KD of < 10 nM at a NaCl concentration of 0.1 M. This dissociation constant is three orders of magnitude smaller than the KD determined for the single high-affinity Mn2+ site in the mHHRz (KD = 4.4 μM). The measurements of catalytic activity have been performed using fluorescent-labeled RNAs. Compared to the mHHRz, the cis tsHHRz cleaves up to 20-fold faster in the presence of Mg2+/Mn2+ ions with no saturation of the cleavage rates at high metal(II) ion concentrations. This is in good agreement with the last investigations on the trans tsHHRz (Nelson et al. 2005). Thus, the much stronger Mn2+ binding and higher cleavage activity were attributed to the interaction between the two external loops of the tsHHRz which reduces the RNA dynamics and traps the Mn2+ in the tightly folded conformation. Intriguingly, according to the EPR studies the binding constants for Mn2+ ions are several orders higher than the concentration of Mn2+ ions required for the catalytic activity (mHHRz: KD = 4.4 ± 0.5 μM and the Mn2+ concentration required to achieve half of the maximum cleavage rate [Mn2+]1/2 = 4.1 ± 0.6 mM respectively). Therefore, strongly bound Mn2+ ions seem to be needed for the folding of the HHRz, whereas weakly bound metal(II) ions are required to achieve full catalytic activity, and may be directly involved in catalysis. A comparison between the Electron Spin Echo Envelope Modulation (ESEEM) and Hyperfine Sublevel Correlation (HYSCORE) spectra of m- and tsHHRz demonstrates that both binding sites in HHRzs are structurally very similar. This suggests that the Mn2+ is located in both ribozymes between the bases A9 and G10.1 of the sheared G•A tandem basepair, as shown previously and in detail for the mHHRz (Vogt and DeRose 1998, Schiemann et al. 2003). However, the hyperfine spectra of the tsHHRz with 15N labeled G10.1 revealed no difference in comparison with the ones with 14N. This leads to an interpretation that the Mn2+ binding sites in both ribozymes are not identical. In addition, aminoglycoside antibiotic neomycin B inhibits the cleavage activity of both despite of the fact that it displaces the high-affinity Mn2+ ion only from the mHHRz. Hence, binding of neomycin B to the m- and the tsHHRzs probably occurs at different sites and neomycin B displaces only loosely bound Me2+ ions from the tsHHRs, whereas in the mHHRz both the high-affinity ion and the weakly bound ions are replaced. Therefore, it cannot be excluded that weakly bound Mg2+/Mn2+ ions, together with looploop interactions, induce a structural rearrangement which brings the high-affinity ion closer to the cleavage site. In the case of the Diels-Alder ribozyme it possesses five Mn2+ binding sites with KD = 0.6 ± 0.2 μM in solution under conditions where it is catalytically active. The competition experiment with Cd2+ allows to distinguish three different types of Mn2+ binding sites in the Diels-Alder ribozyme including inner-sphere monomeric Mn2+, monomeric Mn2+ bound through water-mediated contacts and electronically coupled dimeric Mn2+. Three Mn2+ ions are more strongly bound to the ribozyme via inner-sphere contacts, whereas two other Mn2+ ions form water-mediated outer-sphere contacts with the nucleotides of the ribozyme. The inner-sphere Mn2+ with the highest affinity and the fourth Mn2+ ions added to the ribozyme form a dimer with a Mn2+-Mn2+ distance of ~6 Å (as arises from simulations). Moreover, an addition of the product analog inhibitor (AMDA) to the [Diels-Alder ribozymes/ Mn2+] complex shows no conformational changes in the Mn2+ binding pockets. This is in good agreement with the recent studies which suggest that the Diels-Alder ribozyme is preorganized (Keiper et al. 2004). Some considerations on the evolution of the project (Outlook) There may be several venues of continuation of this project, which exploit on unique combination of EPR experiments and biochemical studies on RNA. This combination may allow us to significantly contribute to understanding of metal role in HHRz catalysis. Since the tsHHRz possesses the high affinity Mn2+ binding site (Kd < 10 nM) it creates a possibility to find conditions where the structural site is occupied by Mn2+, while catalytic sites are occupied by Mg2+ ions. If these conditions will be established by EPR titration, a set of standard biochemical experiments may be designed to look at the kinetic of cleavage and differentiate the “structural” and catalytic effects. The other experiment would be to look at the Mn2+ binding site in the tsHHRz in comparison with P1 and P1/P2 complexes and compare the results with the ones for the mHHRz. No matter the answer, P1 can be used as a simpler model to study the effect of tertiary structure on Mn2+ binding. A set of the tsHHRz mutants can be created to observe the mutations affect on Mn2+ binding sites, Mn2+ affinity and correlate the data with the kinetic analysis. FRET-based kinetic assay with fluorophore pairs on P1 and P2 can be designed for the kinetic experiments. Having this system one will be able to perform kinetic measurements 100-fold faster comparing to standard gel procedures (everything will be done in 96-wells). By manipulating the lengths and the sequence of P2 we most likely will be able to use FRET assay for the chemical step analysis (provided Kd > k2), and measure it using stop-flow system with time resolution of microseconds. And finally, one will be able to quantitatively measure the effect of neomycin B on the tsHHRz. Another interesting possibility would be to look at the state of metal(II) in the tsHHRz – enzyme alone (dissociated product) and in the enzyme-product complex and compare with the full-length tsHHRz. It will provide the information about the local rearrangements upon catalysis and the role of metal(II) ions. Furthermore, additional pulse-EPR experiments using 15N labeling have to be performed in order to reveal the location of the high-affinity Mn2+ binding site in the tsHHRz. Additionally, paramagnetic Mn2+ ions can be localized within the global fold of HHRzs using PELDOR and site-directed spin labeling. Further characterization of the high-affinity binding site in the tsHHRz can be performed using high-field ENDOR measurements in order to obtain the 14N and 31P tensors.
Unlimited self-renewal is an absolute prerequisite for any malignancy, and is the ultimate arbiter of the continuous growth and metastasis of tumors. It has been suggested that the self-renewal properties of a tumor are exclusively contained within a small population, i.e., the so-called cancer stem cells. Enhanced self-renewal potential plays a pivotal role in the development of leukemia. My data have shown that APL associated translocation products PML/RARalpha and PLZF/RARalpha increased the replating efficiency of mouse lin-/Sca1+ hematopoietic stem cells (HSCs). This effect is partly mediated by induction of gamma–catenin which is an important mediator of the Wnt signaling pathway and has been shown to be up regulated by the AML associated translocation products(AATPs). Suppression of gamma–catenin by siRNA can abrogate the increased replating efficiency induced by AATPs. Transduction of gamma–catenin in lin-/Sca1+ HSCs led to increased replating efficiency and the expression of stem cell markers Sca1 and c-kit. Additionally it induced accelerated cell cycle progression of mouse bone marrow HSCs. Transduction/transplantation mouse models have shown that ectopic expression of gamma–catenin in HSCs led to acute myeloid leukemia without maturation. These data suggest important roles of Wnt signaling pathway in the leukemogenesis induced by PML/RARalpha, PLZF/RARalpha and AML1/ETO. In contrast to AATPs, CML and Ph+-ALL associated translocation products p185(BCR-ABL) and p210(BCR-ABL) did not affect the self-renewal potential of hematopoietic stem/progenitor cells. However my studies indicated that their reciprocal translocation products p40(ABL/BCR) and p96(ABL/BCR) actually increased the replating efficiency of hematopoietic stem/progenitor cells. The effect is stronger when induced by p96(ABL/BCR) than by p40(ABL/BCR). It is very intriguing that p96(ABL/BCR) can activate Wnt signaling and up regulate the expression of HoxB4. Transduction/transplantation mouse model has shown that p40(ABL/BCR) and p96(ABL/BCR) both have their own leukemogenic potential. Given the fact that leukemic stem cells maintain the growth of tumor and are the origin of relapse, the cure of leukemia is dependent on the eradication of the leukemic stem cell and abrogation of aberrantly regulated self-renewal capability. Both t-RA and As2O3 have been shown to induce complete remission in APL patients with PML/RARalpha translocation product. However, t-RA as a single agent achieves completeremission (CR) but not complete molecular remissions (CMR). Therefore, virtually all patients will experience a relapse within a few months. In contrast to t-RA, As2O3 as a single agent is able to induce CR as well as CMR followed by long-term relapse-free survival in about 50% of APL patients even if relapsed after treatment with t-RA-containing chemotherapy regimens. Nothing is known about the mechanisms leading to the complete different clinical outcomes by the two compounds although both have been shown to induce differentiation of blast cells, proliferation arrest, induction of apoptosis and degradation of PML/RARalpha. We investigated the effect of t-RA and arsenic on PML/RARalpha-expressing cell population with stem cell capacity derived from the APL cell line NB4 as well as Sca1+/lin- murine bone marrow cells. We found that t-RA did not reduce the replating efficiency in PML/RARalpha- and PLZF/RARalpha-infected Sca1+/lincells whereas it selected small compact colonies representing very early progenitor cells. T-RA was unable to reduce the capacity to form colony forming units-spleen (CFU-S) of Sca1+/lin-cells expressing PML/RARalpha, additionally t-RA did not impair the capability of engraftment of NB4 cells in NOD/SCID mouse. On the contrary to t-RA, As2O3 abolished the aberrant self-renewal potential of Sca1+/lin- cells expressing PML/RARalpha. As2O3 not only abolished the replating efficiency of PML/RARalpha positive cells but also completely abrogated the ability of PML/RARalpha-positive HSC to produce CFU-S in vivo. On the contrary to As2O3, t-RA increased the absolute cell number and the percentage of cells in the side population with respect to the whole cell population in NB4 cells. Taken together these data suggest that arsenic but not all-trans retinoic acid overcomes the aberrant stem cell capacity of PML/RARalpha positive leukemic stem cells. My data prove for the first time that there is a direct relationship between the capacity of compounds to effectively target the LSC and their capacity to eradicate the leukemia, and, thereby, to induce complete molecular remission and long-term relapse-free survival. Thus, in order to increase the curative potential of leukemia therapies, future studies need to include the effect of given compounds on the stem cell compartment to determine their ability to eradicate the LSC.
Oxidative stress attenuates the NO-cGMP pathway, e.g. in the vascular system, through scavenging of free NO radicals by superoxide O2•-, by inactivation of soluble guanylyl cyclase (sGC) via oxidation of its central Fe2+ ion, and by down-regulation of sGC protein levels. While the former pathways are well established, the molecular mechanisms underlying the latter are still obscure. Using oxidative sGC inhibitor ODQ we demonstrate rapid down-regulation of sGC protein in mammalian cells. Co-incubation with proteasomal inhibitor MG132 results in accumulation of ubiquitinated sGC whereas sGC activator BAY 58–2667 prevents ubiquitination. ODQ-induced down-regulation of sGC is mediated through selective ubiquitination of its b subunit, and BAY 58–2667 abrogates this effect. Ubiquitination of sGC-b is dramatically enhanced by E3 ligase CHIP. Our data indicate that oxidative stress promotes ubiquitination of sGC b subunit through E3 ligase CHIP, and that sGC activator 58–2667 reverts this effect, most likely through stabilization of the heme-free b subunit. Thus the deleterious effects of oxidative stress can be counter-balanced by an activator of a key enzyme of vascular homeostasis.
To facilitate the measurement of intramolecular distances in solvated RNA systems, a combination of spin-labeling, electron paramagnetic resonance (EPR), and molecular dynamics (MD) simulation is presented. The fairly rigid spin label 2,2,5,5-tetramethyl-pyrrolin-1-yloxyl-3-acetylene (TPA) was base and site specifically introduced into RNA through a Sonogashira palladium catalyzed crosscoupling on column. For this purpose 5-iodouridine, 5-iodo-cytidine and 2-iodo-adenosine phosphoramidites were synthesized and incorporated into RNA-sequences. Application of the recently developed ACE (R) chemistry presented the main advantage to limit the reduction of the nitroxide to an amine during the oligonucleotide automated synthesis and thus to increase substantially the reliability of the synthesis and the yield of labeled oligonucleotides. 4-Pulse Electron Double Resonance (PELDOR) was then successfully used to measure the intramolecular spin–spin distances in six doubly labeled RNA-duplexes. Comparison of these results with our previous work on DNA showed that A- and B-Form can be differentiated. Using an all-atom force field with explicit solvent, MD simulations gave results in good agreement with the measured distances and indicated that the RNA A-Form was conserved despite a local destabilization effect of the nitroxide label. The applicability of the method to more complex biological systems is discussed.
Riboswitches are highly structured elements in the 50-untranslated regions (50-UTRs) of messenger RNA that control gene expression by specifically binding to small metabolite molecules. They consist of an aptamer domain responsible for ligand binding and an expression platform. Ligand binding in the aptamer domain leads to conformational changes in the expression platform that result in transcription termination or abolish ribosome binding. The guanine riboswitch binds with high-specificity to guanine and hypoxanthine and is among the smallest riboswitches described so far. The X-ray-structure of its aptamer domain in complex with guanine/ hypoxanthine reveals an intricate RNA-fold consisting of a three-helix junction stabilized by longrange base pairing interactions. We analyzed the conformational transitions of the aptamer domain induced by binding of hypoxanthine using highresolution NMR-spectroscopy in solution. We found that the long-range base pairing interactions are already present in the free RNA and preorganize its global fold. The ligand binding core region is lacking hydrogen bonding interactions and therefore likely to be unstructured in the absence of ligand. Mg2+-ions are not essential for ligand binding and do not change the structure of the RNA-ligand complex but stabilize the structure at elevated temperatures. We identified a mutant RNA where the long-range base pairing interactions are disrupted in the free form of the RNA but form upon ligand binding in an Mg2+-dependent fashion. The tertiary interaction motif is stable outside the riboswitch context.
Membranes are essential for life, because a cell must separate itself from the environment to keep its molecules from dissipating away and also must keep out foreign molecules that disturb them or their cell components. However, the cell must communicate with the environment and adapt to the external conditions, needs to pump in nutrients and release toxic products of its metabolism. Membrane proteins present in the membranes of the cell and cell organelles, help the cell to gather information about the environment and perform various biological processes. Membrane proteins perform a wide range of biological functions including respiration, signal transduction and transport. Despite their high importance in biological function, only few structures have been determined because of the difficulties in producing high amounts of membrane proteins and obtaining good quality crystals. This Ph. D. thesis involves the study of different kinds of cytochrome oxidases and a membrane anchored cytochrome oxidase electron donor. Though structures of many cytochrome oxidases are known to date, there exist many different types of oxidases in different organisms, which help the organism to survive under unfavorable environmental conditions. The structural differences between these terminal oxidases which make the organism to survive in extreme environments are unclear. To investigate these, structures of different types of oxidases are necessary. Therefore, we are interested in revealing the structural details of different types of oxidases. The different types of oxidase I worked with were the caa3 HiPIP:oxygen oxidoreductase from Rhodothermus marinus, the aa3-type quinol oxidase from Acidianus ambivalens and bd-type quinol oxidase from three different organisms (Escherichia coli, Bacillus thermodenitrificans and Aquifex aeolicus). Besides the protein from E. coli all other proteins are from thermophilic organisms from which the proteins obtained are generally believed to be highly stable. The presence of a high content of charged amino acids that enhances the occurrence of salt bridges contributes to the stability of thermophilic proteins. ....
