Seit der TIMSS- und PISA-Studie sind sie wieder einmal Thema der bildungspolitischen Diskussion: die naturwissenschaftlichen Fächer in unseren Schulen. Deutschen Schülerinnen und Schülern wird bescheinigt: Sie haben nur mangelnde Kenntnisse, verstehen zu wenig, sind nicht recht in der Lage, Fragestellungen methodisch anzugehen, und denken zu wenig darüber nach, wie sie naturwissenschaftliche Probleme lösen könnten. Aber auch viele Erwachsene geben offen zu, besonders von den »harten« Naturwissenschaften wie Chemie wenig zu verstehen und sich nie besonders dafür interessiert zu haben. Wie kommt es, dass eine Wissenschaft, die wesentlich zum Verständnis unserer stofflichen Umwelt beiträgt und deren praktische Anwendung unser tägliches Leben in hohem Maße beeinflusst, auf ein so geringes Interesse stößt?
Bei der Expression einer Phospholipase A2 aus Soja in Aspergillus oryzae (Probe PL-1007), Trichoderma viride (Probe PL-1008) und Pichia pastoris (Probe PL-1035) wurde neben der erwarteten Phospholipaseaktivität auch eine deutliche Lysophospholipaseaktivität beobachtet. Eine Trennung dieser Aktivitäten war nur mittels einer Free-Flow-Elektrophorese kurzzeitig möglich, bevor sich in jeder Fraktion wieder das ursprüngliche Aktivitätsverhältnis zwischen Phospholipase und Lysophospholipase einstellte. Zur näheren Untersuchung und Charakterisierung dieser Enzymsysteme wurden für einen gaschromatischen Aktivitätstest hochspezifisch substituierte Substrate eingesetzt, die eine parallele Bestimmung der Phospholipase- und Lysophospholipaseaktivitäten ermöglichten. So konnte gezeigt werden, dass nicht in allen Organismen eine Phospholipase A2 exprimiert wird, sondern es in Pichia pastoris (Probe PL-1035) zur Expression einer Phospholipase A1 kommt. Im Falle der Probe PL-1007 konnte die beobachtete Lysophospholipaseaktivität durch Versuche mit veränderten Substratverhältnissen zwischen Lysophospholipase- und Phospholipasesubstrat einem separaten aktiven Zentrum zugeschrieben werden. Durch elektrophoretische Trennungsversuche konnte gezeigt werden, dass es sich nicht nur um separate aktive Zentren, sondern um verschiedene Enzyme handelt. Die Tatsache, dass sich das Aktivitätsverhältnis nach der Trennung selbständig wieder einstellt, lässt das Vorliegen von Faltungsisomeren vermuten. Bezüglich ihrer katalytischen Eigenschaften weisen alle drei Enzymsysteme eine große Ähnlichkeit auf. Sowohl die Phospholipase- wie auch die Lysophospholipaseaktivitäten sind erst ab einer Reaktionstemperatur von über 70°C nicht mehr nachweisbar. Das Aktivitätsmaximum wurde in allen drei Fallen zwischen 45°C und 55°C beobachtet. Auch die pH-Bereiche in denen eine maximale enzymatische Aktivität zu beobachten ist, liegen mit pH 3,6 (PL-1007) bis pH 4,3 (PL-1035) für die Phospholipaseaktivitäten und pH 4,3 (PL-1007 / PL-1008) und pH 4,6 (PL-1035) in ähnlichen Bereichen. Die Aktivität der untersuchten Enzymsysteme zeigt jedoch im beobachteten pH-Bereich von pH 3 bis pH 5 nur eine geringe pH-Abhängigkeit. Deutlichere Unterschiede konnten jedoch für die Substratspezifitäten nachgewiesen werden. Die Phospholipasen A2 zeigen tendenziell höhere Umsätze bei Substraten mit C 12:0 bis C 16:0 Fettsäureresten, während die Phospholipase A1 aus Probe PL-1035 maximale Umsätze bei C 18:0 Fettsäureresten aufweist. Bei allen Enzymproben jedoch bleiben die Umsatzraten der ein- bis mehrfach ungesättigten Fettsäurereste hinter denen der gesättigten zurück. Lediglich bei der Probe PL-1007 sind die Aktivitätsunterschiede zwischen gesättigten und ungesättigten Substraten nicht signifikant. Neben der Untersuchung der katalytischen Eigenschaften konnte im Rahmen dieser Arbeit die in der Literatur mehrfach aufgestellte These der Hemmung der Phospholipaseaktivität in Gegenwart von Uteroglobin bestätigt werden. Ein Hemmungsmechanismus aufgrund direkter Wechselwirkung der Enzyme konnte ausgeschlossen werden. Vielmehr konnte zweifelsfrei bewiesen werden, dass der Hemmungsmechanismus bei den hier eingesetzten Enzymsystemen auf einer Bindung des für die Phospholipasereaktion essentiellen Ca2+ durch das Uteroglobin zurückzuführen ist.
Rhodopsin-based voltage imaging tools for use in muscles and neurons of Caenorhabditis elegans

Genetically encoded voltage indicators (GEVIs) based on microbial rhodopsins utilize the voltage-sensitive fluorescence of all-trans retinal (ATR), while in electrochromic FRET (eFRET) sensors, donor fluorescence drops when the rhodopsin acts as depolarization-sensitive acceptor. In recent years, such tools have become widely used in mammalian cells but are less commonly used in invertebrate systems, mostly due to low fluorescence yields. We systematically assessed Arch(D95N), Archon, QuasAr, and the eFRET sensors MacQ-mCitrine and QuasAr-mOrange, in the nematode Caenorhabditis elegans ATR-bearing rhodopsins reported on voltage changes in body wall muscles (BWMs), in the pharynx, the feeding organ [where Arch(D95N) showed approximately 128% ΔF/F increase per 100 mV], and in neurons, integrating circuit activity. ATR fluorescence is very dim, yet, using the retinal analog dimethylaminoretinal, it was boosted 250-fold. eFRET sensors provided sensitivities of 45 to 78% ΔF/F per 100 mV, induced by BWM action potentials, and in pharyngeal muscle, measured in simultaneous optical and sharp electrode recordings, MacQ-mCitrine showed approximately 20% ΔF/F per 100 mV. All sensors reported differences in muscle depolarization induced by a voltage-gated Ca2+-channel mutant. Optogenetically evoked de- or hyperpolarization of motor neurons increased or eliminated action potential activity and caused a rise or drop in BWM sensor fluorescence. Finally, we analyzed voltage dynamics across the entire pharynx, showing uniform depolarization but compartmentalized repolarization of anterior and posterior parts. Our work establishes all-optical, noninvasive electrophysiology in live, intact C. elegans.
Die vorliegende Arbeit beschäftigt sich mit unterschiedlichen Aspekten der Faltung von Tendamistat, einem aus 74 Aminosäuren bestehenden a-Amylase lnhibitor aus Streptomyces tendae. Bei der oxidativen Rückfaltung des Tendamistats konnten bisher drei kinetische Phasen (t1, t2 und t3) identifiziert werden. Die mittlere (t2) bzw. langsame (ti) kinetische Phase, resultiert als Folge einer cis/trans lsomerisierung um eine der drei Xaa-Pro Bindungen im Tendamistat. Wie im ersten Teil dieser Arbeit gezeigt werden konnte, entsteht die prolinabhängige lsomerisierungsreaktion während der Rückfaltung (t3) als Folge einer trans -> cis Umlagerung um die A1a49-Pro50 Amidbindung nach Entfaltung des Proteins. Der Nachweis einer solchen lsomerisierung gelang auf rekombinantem Weg, durch gezielten Austausch der einzelnen Proline. Im Gegensatz dazu resultiert die mittlere kinetischen Phase (t2) als Folge einer Prolin-unabhängigen cis/trans-lsomerisierungsreaktion um mindestens eine vorhandenen nicht-Prolinbindungen im Protein. Diese Schlußfolgerung konnte durch Vergleich mit zahlreichen Literaturdaten und Experimenten weiter gestützt werden. Tendamistat ist damit das erste Protein in der Literatur, bei dem es gelang, eine solche Prolin-unabhängige lsomerisierungsreaktion experimentell nachzuweisen. Wie mit Hilfe von weiteren Substitutionsexperimenten im zweiten Teil der Arbeit gezeigt werden konnte, tragen hydrophobe Oberflächencluster entscheidend zur Stabilisierung des lnhibitors bei. Die dabei gemessenen Stabilisierungsbeiträge stehen in einem linearen Verhältnis zum Seitenkettenvolumen der eingeführten Aminosäurereste (Alanin << Valin << Isoleucin). Hierdurch ließ sich zeigen, daß der stabilisierende Einfluß solcher Cluster im Wesentlichen auf das Vorhandensein von hydrophoben Wechselwirkungen zurückzuführen ist. Für die Faltung von Proteinen, die wie das Tendamistat ausschließlich aus ß-Faltblättern bestehen, spielt die Bildung von ß-Hairpinstrukturen eine entscheidende Rolle Die in dieser Arbeit untersuchten ß-Hairpinmutanten L14A bzw. N25A zeigen im Vergleich zum Wildtyp eine extreme Destabilisierung der ß-Faltblattstruktur. Durch Vergleich der Aktivierungsenergien (DeltadeltaGD-TS) mit den freien Enthalpien (DeltadeltaG0N-D) konnte die strukturelle Entwicklung des ß-Hairpins im Übergangszustand an den beiden Positionen 14 bzw. 25 mitverfolgt werden (~-Wert Analyse). Entgegen den Erwartungen zeigen diese Untersuchungen, daß beide Positionen im Übergangszustand noch weitestgehend solvatisiert vorliegen. Damit scheint die nativ-ähnliche Zusammenlagerung des ersten ß-Hairpins erst nach Erreichen des Übergangszustands stattzufinden. Alle in dieser Arbeit vorgelegten Daten über die Faltung des alpha-Amylase lnhibitors sind konsistent mit dem Nukleations-Kondensations Modell. Analog der Tröpfchenbildung bei unterkühlten Gasen kommt es nach dem kollabieren der Proteinkette als Reaktion auf die veränderten Lösungsmittelbedingungen zur intramolekularen Suche nach Nukleationsstellen. Der Übergangszustand wird erreicht, wenn sich eine kritische Zahl stabilisierender Kontakte (unspezifische Nukleation) ausbilden konnte. Ob an einem solchen Nukleationskern stets bestimmte Seitenketten beteiligt sind (spezifische Nukleation), bleibt an dieser Stelle noch offen.
Paläobotanische Untersuchungen an Euramerischen Kohlenbecken haben an der Westfal/Stefan-Grenze in früheren Arbeiten einen deutlichen, weitgehend klimatisch gesteuerten Florenwechsel erkennen lassen. Desweiteren wurden in Kohlen aus dem Saar/Nahe-Becken beginnend mit dem obersten Westfal D erstmals Diageneseprodukte von Isoarborinol bzw. Fernen/Fernenol nachgewiesen, für die Koniferen, Cordaiten oder Farnsamer als mögliche Bioproduzenten vorgeschlagen wurden. Im Rahmen der vorliegenden Arbeit konnten die Arboran-/Fernanderivate MAPH, MATH, DAPH 1 und DAPH 2 in den Gesamtextrakten von Kohlen und Sedimenten aus dem Stefan des Saar/Nahe-Beckens durchgängig identifiziert werden, während die Verbindungen im Westfal lediglich in Proben aus dem obersten Westfal D auftraten. Folglich sind die Arboran-/Fernanderivate tatsächlich in besonderem Maße dazu geeignet, den Florenwechsel an der Westfal/Stefan-Grenze auf molekularer Basis zu beschreiben. Um einzugrenzen, zu welcher Pflanzengruppe die Bioproduzenten der Ausgangsverbindungen der Arboran-/Fernanderivate gehören, wurden isolierte Makrofossilien verschiedener Pflanzengruppen aus verschiedenen Euramerischen Kohlenbecken organisch-geochemisch analysiert. Dabei konnten MATH, MAPH, DAPH 1 und DAPH 2 in nahezu allen Gesamtextrakten fossiler Cordaiten-Reste identifiziert werden. In den Extrakten von Sediment-Vergleichsproben, die in unmittelbarer Nähe der Cordaiten-Reste entnommen wurden, konnten die Verbindungen dagegen nicht bzw. nur in vergleichsweise geringen Konzentrationen identifiziert werden. Ebenso waren die Arboran-/Fernanderivate in den Gesamtextrakten fossiler Koniferen-Reste sowie in den Extrakten verschiedener Farnsamerarten (Alethopteris, Dicroidium, Lescuropteris, Macroneuropteris, Neuropteris) nicht enthalten. Lediglich in der extrahierbaren organischen Substanz einiger fossiler Odontopteris-Reste aus dem Blanzy-Montceau-Becken (Frankreich) konnten MAPH und MATH (sowie teilweise DAPH 1 und DAPH 2) identifiziert werden. Allerdings ist das Auftreten der Verbindungen in diesen Odontopteris-Extrakten wahrscheinlich auf eine Überprägung des Pflanzenmaterials durch das umgebende Sediment zurückzuführen, da die Verbindungen in den Sediment-Vergleichsproben in höheren bzw. ähnlichen Konzentrationen enthalten sind. Insgesamt sind daher in den oberkarbonischen Kohlenbecken die Cordaiten als einer, möglicherweise sogar als „die“ Bioproduzenten der Ausgangsverbindungen der Arboran-/Fernanderivate MATH, MAPH, DAPH 1 und DAPH 2 anzusehen. Die deutlich negativere Kohlenstoffisotopie (-31,68 ‰) einer Sedimentprobe aus der Bohrung Wemmetsweiler-Nord, die gleichzeitig die höchsten Arboran-/Fernanderivat-Konzentrationen enthält, weist auf eine verstärkte mikrobielle Überarbeitung des organischen Materials hin. Fluoreszenz- und Auflichtmikroskopie-Untersuchungen zeigen zudem einen erheblichen, ebenfalls auf bakterielle Aktivität hindeutenden Bituminitanteil, während Relikte von höheren Landpflanzen nur geringfügig vertreten sind. Dies legt den Schluß nahe, daß die Arboran-/Fernanderivate in dieser Probe nicht von Cordaiten, sondern alternativ von Bakterien (oder Algen) abstammen. In diesem Fall ist Isoarborinol als biologischer Vorläufer anzunehmen und es tritt eine deutliche Verschiebung der Kohlenstoffisotopie-Werte zu negativeren Werten auf. Bei den Cordaiten ist eine derartige Isotopenverschiebung dagegen nicht zu beobachten, so daß für MATH, MAPH, DAPH 1 und DAPH 2 Fernen bzw. Fernenol als biologische Vorläufer anzunehmen sind.
Die Sonne strahlt weltweit pro Tag genügend Licht ein, um den Weltenergiebedarf für ein ganzes Jahr abzudecken. Somit ist sie die Quelle aller erneuerbarer Energien, denn neben der Erzeugung von Elektrizität aus Licht (Photovoltaik) regelt sie die Gezeiten und damit auch Wind und Wellen, die bei der Windkraft und in Gezeitenkraftwerken genutzt werden. Außerdem liefert sie die Energie für die Photosynthese in nachwachsenden Rohstoffen. Es gibt diesbezüglich nur ein grundlegendes Problem: Erneuerbare Energien fi nden wir in ausreichender Menge vor allem an Stellen mit mangelnder Infrastruktur. Sonnenenergie gibt es am meisten in der Wüste, Wind auf dem Meer und Biomasse im Dschungel. An Orten hoher Industrialisierung und damit auch hoher Bevölkerungsdichte ist für die »Erneuerbaren « so gut wie kein Platz. Es gibt demnach kein Energieproblem, aber ein Problem der Energiespeicherung und des Energietransportes.
Weltweit nehmen Kohle-, Öl- und Erdgasvorräte ab, der Energiebedarf dagegen steigt dramatisch an. Regenerative Energien mindern zwar die steigenden Klimagefahren, können aber unseren zukünftigen Energiebedarf in Ballungszentren kaum decken. Nach Einschätzung zahlreicher Experten gehört dem Wasserstoff die Zukunft. Er wird aber derzeit nahezu ausschließlich aus fossilen Brennstoffen gewonnen; damit bleibt auch diese Ressource endlich - ganz zu schweigen von ihrem hohen Gefährdungspotenzial. - Das neuartige Energiekonzept einer solaren und damit kohlenstoffunabhängigen Wasserstoffwirtschaft bedarf zur technischen Realisierung eines Zwischenspeichers für regenerative Energien. Dieser zukünftige Energieträger sollte synthetisch einfach erzeugbar sein, in unbegrenztem Maß zur Verfügung stehen oder zumindest recycelbar sein, die Energie permanent speichern und gefahrlos transportierbar sein, eine hohe Energiedichte aufweisen und kein Kohlendioxid oder andere (Klima-) Schadstoffe freisetzen. - Das Element Silicium kann zu einem maßgeschneiderten Bindeglied zur Ankoppelung dezentraler regenerativer Energieerzeugung an eine ebenso dezentrale Wasserstoffwirtschaft an jedem beliebigen Ort werden. Der Transport und die Speicherung von Silicium sind - im Gegensatz zu Öl oder besonders zu Wasserstoff - ohne Gefährdungspotenzial und/oder hohe Energieverluste möglich und erfordern nur eine technische Infrastruktur, wie sie auch für Kohle benötigt wird.
Global reserves of coal, oil and natural gas are diminishing; global energy requirements however are dramatically increasing. Renewable energy sources lower the threat to the earth’s climate but are not able to meet the energy consumption in major urban areas. The opinion of many experts is that the future will be dominated by hydrogen. However, this gas is essentially totally manufactured from fossil fuels and is hence of limited abundance – not to mention the hazards involved in its utilisation. - A novel energy concept involving solar and thus carbon-independent hydrogen-based technology necessitates an intermediate storage vehicle for renewable energy. This future energy carrier should be simple to manufacture, be available to an unlimited degree or at least be suitable for recycling, be able to store and transport the energy without hazards, demonstrate a high energy density and release no carbon dioxide or other climatically detrimental substances. - Silicon successfully functions as a tailor-made intermediate linking decentrally operating renewable energy-generation technology with equally decentrally organised hydrogen-based infrastructure at any location of choice. In contrast to oil and in particular hydrogen, the transport and storage of silicon are free from potential hazards and require a simple infrastructure similar to that needed for coal.
The Na+-F1F0-ATPase operon ofAcetobacterium woodii was recently shown to contain, among eleven atp genes, those genes that encode subunita and b, a gene encoding a 16-kDa proteolipid (subunit c 1), and two genes encoding 8-kDa proteolipids (subunits c 2 andc 3). Because subunits a,b, and c 1 were not found in previous enzyme preparations, we re-determined the subunit composition of the enzyme. The genes were overproduced, and specific antibodies were raised. Western blots revealed that subunits a,b, and c 1 are produced and localized in the cytoplasmic membrane. Membrane protein complexes were solubilized by dodecylmaltoside and separated by blue native-polyacrylamide gel electrophoresis, and the ATPase subunits were resolved by SDS-polyacrylamide gel electrophoresis. N-terminal sequence analyses revealed the presence of subunitsa, c 2, c 3,b, δ, α, γ, β, and ε. Biochemical and immunological analyses revealed that subunitsc 1, c 2, andc 3 are all part of the c-oligomer, the first of a F1F0-ATPase that contains 8- and 16-kDa proteolipids.
