Untersuchung der Wirkung von Cyclooxygenase-Inhibitoren auf die Entstehung der Arteriosklerose

In der vorliegenden Arbeit wurde der Einfluss der COX-2-selektiven NSAIDs Celecoxib und Rofecoxib sowie des traditionellen nichtselektiven NSAIDs Naproxen auf die Entstehung und Entwicklung von Arteriosklerose in ApoE-defizienten Mäusen untersucht. Befunde über kardiovaskuläre Risiken dieser Medikamentengruppe sowie die Beobachtung, dass COX-2 in arteriosklerotischen Plaques exprimiert wird, ließen sowohl die Hypothese von pro- als auch von antiatherogenen Wirkungen zu. Zunächst wurde die Funktionsfähigkeit des Versuchsdesigns nachgewiesen. In den Aorten von Diätgefütterten ApoE-defizienten Mäusen entwickelten sich im Alter von 22 Wochen Plaques, in denen typische atherogene sowie proinflammatorische Proteine exprimiert wurden. Die Effektivität und Selektivität der jeweiligen NSAIDs wurde durch Messung der Urinkonzentration relevanter Metabolite nachgewiesen, die aus der COX-2-vermittelten PGI2- und der COX-1-vermittelten TXA2-Synthese stammten. Erwartungsgemäß hemmten sowohl die Coxibe als auch Naproxen die Synthese von PGI2, während die TXA2-Produktion nur unter Naproxen reduziert wurde. Im Vergleich zu Kontrolltieren führte die chronische Behandlung mit Celecoxib und Rofecoxib tendenziell zu kleineren Plaques in der Aorta von diätgefütterten ApoE-defizienten Mäusen. In den Plaques wurde die Proteinexpression von COX-2 durch die Behandlung mit den NSAIDs nicht beeinflusst. Dies deutet darauf hin, dass die Reduktion der Plaques in den Aorten eher durch eine Hemmung der COX-2-Aktivität oder durch COX-unabhängige Effekte als über eine Inhibition der COX-2-Proteinexpression zustande kommt. Die Blutfettwerte wurden durch die eingesetzten NSAIDs nicht verändert, was den Daten aus der Literatur entspricht. ApoE-defiziente Mäuse, die normales Futter erhalten hatten und mit Celecoxib oder Rofecoxib behandelt wurden, entwickelten vereinzelt intimale Läsionen, während Kontrolltiere oder Naproxen-behandelte Mäuse in keinem Fall Plaques zeigten. Diese Ergebnisse könnten einen Hinweis dafür liefern, dass durch Coxibe COX-unabhängige Effekte ausgelöst werden, welche die Arteriogenese einleiten. Die umfangreichen Untersuchungen lassen auf eine unterschiedliche Beteiligung von NSAIDs bei der Arteriogenese schließen. Dabei scheinen die selektiven COX-2-Inhibitoren Celecoxib und Rofecoxib eine andere Rolle als Naproxen, dem nichtselektiven COX-2-Inhibitor, zu spielen. In ApoEdefizienten Diät-Mäusen wurde die Arteriosklerose durch Celecoxib und Rofecoxib über COX-2-abhängige Mechanismen inhibiert, wobei der Effekt vermutlich durch den stärker ausgeprägten Effekt der Hypercholesterinämie überlagert war. In den Kontroll-Mäusen (mit Normalfutter) dagegen könnte die Arteriogenese unter Celecoxib und Rofecoxib durch COX-unabhängige Mechanismen eingeleitet worden sein. Dabei scheint das Stadium der Arteriosklerose eine wichtige Rolle zu spielen. Zusammenfassend deuten die Ergebnisse daraufhin, dass Coxibe die Initiation der Arteriosklerose eher begünstigen, während bei weiter fortgeschrittenen Plaques tendenziell antiatherogene Effekte zu beobachten waren, die möglicherweise auf die entzündungshemmenden Eigenschaften der Substanzen zurückzuführen sind. Nachdem jedoch diese Effekte bisher nur in Maus-Modellen beobachtet und untersucht wurden, bleibt die Frage, wie NSAIDs thrombotische Ereignisse im Patienten auslösen, weiterhin offen. Erst kürzlich wurde beschrieben, dass unter Rofecoxib schon nach kurzer Behandlungsdauer Herzinfarkte ausgelöst werden können (Levesque et al. 2006). Weitere Daten deuten daraufhin, dass NSAIDs über COX-unabhängige Mechanismen dazu führen, dass sogar nach dem Absetzen des Medikament auf lange Sicht ein erhöhtes Risiko für die Entwicklung kardiovaskuläre Erkrankungen bestehen bleibt (Merck, APPROVe Report 2006; unter www.merck.com). Die genauen Mechanismen dieser Langzeiteffekte von NSAIDs auf das vaskuläre System sind zurzeit nach wie vor unklar. Abschließend kann jedoch aufgrund der aktuellen Datenlage vermutet werden, dass Coxibe eine Schädigung von vaskulären Zellen verursachen können und das Risiko für kardiovaskuläre Effekte eher erhöhen als antiatherogene Effekte zu erzeugen und die Arteriosklerose zu verbessern.
Das Enzym 5-Lipoxygenase (5-LO) spielt eine essentielle Rolle in der Biosynthese der Leukotriene, bioaktiver Metabolite der Arachidonsäure (AA), die an einer Vielzahl entzündlicher und allergischer Erkrankungen beteiligt sind. Die 5-LO wird bevorzugt in Zellen myeloiden Ursprungs wie Granulozyten, Monozyten oder B-Lymphozyten exprimiert. In die Regulation der zellulären 5-LO-Aktivität in der Epstein-Barr Virus-transformierten B-lymphozytären Zelllinie BL41-E95-A sind Caspasen, Aspartat-spezifische Cysteinproteasen, involviert. Das Passagieren von BL41-E95-A führt zu einer Erhöhung der Proliferationsrate der B-Lymphozyten sowie zu einem deutlichen Verlust der 5-LO-Aktivität, der mit dem Auftreten eines 62 kDa-Spaltproduktes der 5-LO und einer signifikanten Aktivitätserhöhung der Caspase-8 und -6 korreliert. Isolierte humane 5-LO wird durch rekombinante Caspase-6 zwischen Asp170 und Ser171 zu einem 58 kDa-Fragment in vitro gespalten, wobei das Tetrapeptid VEID170 innerhalb der 5-LO als Erkennungsmotiv für den Angriff der Caspase-6 dient. In einigen weiteren untersuchten Zelllinien wie Mono Mac 6 (MM6), RBL-1, PMNL oder HeLa, die nicht den B-Lymphozyten angehören, konnte die 5-LO-Spaltung weder durch das Passagieren von Zellen noch durch die Behandlung mit diversen proapoptotischen Agentien ausgelöst werden. Laut Ergebnissen aus in vitro-Untersuchungen scheinen 5-LO-positive HeLa- bzw. MM6-Zellen einen Faktor zu exprimieren, der die 5-LO direkt oder indirekt vor dem Angriff der Caspase-6 und anschließender Prozessierung schützt. Die in den BL41-E95-A-Zellen beobachtete Aktivierung der Caspasen mit anschließender Prozessierung der 5-LO lässt sich durch zwei Pflanzeninhaltsstoffe supprimieren, das Hyperforin (HP) aus Johanniskraut-Extrakten und das Myrtucommulon (MC) aus Myrte-Blättern. Beide Verbindungen scheinen in B-Lymphozyten zu einer Hemmung der Caspasen-Aktivierung zu führen. Nichtsdestotrotz führt die Behandlung der B-Lymphozyten mit HP bzw. MC zu einem apoptotischen Tod der Zellen. Offensichtlich wird dabei ein (unbekannter) einzigartiger Mechanismus der Apoptose-Induktion ausgelöst. In der vorliegenden Arbeit konnte zum ersten Mal eine potente Apoptose-induzierende Wirkung des natürlich vorkommenden Myrtucommulons auf Krebszelllinien gezeigt werden. In allen getesteten Krebszelllinien führte Myrtucommulon zum Zelltod, wobei die HL-60-Zellen mit einem IC50-Wert von 3,26 ± 0,51 µM MC am sensitivsten gegenüber MC-Einfluss waren. Zusätzlich konnte in HL-60- und MM6-Zellen nach MC-Behandlung neben einer erhöhten Caspasen-Aktivität und PARP-Spaltung ein signifikanter DNA-Abbau detektiert werden. Von besonderer Bedeutung ist die Tatsache, dass die zytotoxische MC-Wirkung eine bemerkenswerte Selektivität für entartete Zelllinien zu besitzen scheint und gegenüber nicht-transfizierten Zellen minimal ist.
5-LO is the key enzyme in the biosynthesis of proinflammatory leukotrienes, converting arachidonic acid to 5-HPETE, and in a second step 5-HPETE to leukotriene A4. Although the 5-LO promoter possesses characteristics of so called housekeeping genes, such as lack of TATA/CCAAT boxes and existence of several Sp1 binding sites, the 5 -LO gene is tissue specifically expressed in primarily immune competent cells of myeloid origin including granulocytes, monocytes, macrophages, mast cells and B-lymphocytes. 5-LO gene expression in MM6 and HL-60 cells is strongly induced after differentiation of the cells with TGF-beta and 1,25(OH)2D3. In some monocytic cancer cell lines, such as HL-60 TB and U937, TGF-beta and 1,25(OH)2D3 treatment are not able to activate 5-LO gene transcription. It was demonstrated, that in these cell lines the 5-LO core promoter is heavily methylated and that only demethylation by the DNA methyltransferase inhibitor 5-aza-2 deoxycytidine (Adc) upregulated the 5-LO mRNA levels. It was also shown that the histone deacetylase inhibitor TsA could induce 5-LO mRNA levels, but only in 1,25(OH)2D3/TGF-beta inducible MM6 cells. Interestingly the 1,25(OH)2D3/TGF-beta effect on 5-LO expression is reduced, when combined with TsA. Reporter gene assays revealed that 5-LO promoter activity is strongly induced after 24 h treatment with 330 nM TsA (construct N10 up to 35 fold in HeLa cells). The effect is dependent on the presence of the proximal Sp1 binding site GC4 (-53 bp to –48 bp in relation to the major TIS) in both HeLa and MM6 cells. In vitro binding of the transcription factor Sp1 to this site has been demonstrated in gel shift assays and DNase I footprints. Mutation of the binding site resulted in a loss of basal promoter activity in both 5-LO negative HeLa cells and in 5-LO positive MM6 cells, as well as in the loss of TsA inducibility. The mutational study of different Sp1 binding sites in a larger promoter context revealed the interaction or respectively the additive effect of the multiple Sp1 binding sites of the 5-LO promoter on basal as well as on TsA upregulated promoter activity. However, GC4 seems to be of special relevance for both the basal promoter activity, possibly recruiting the basal transcription machinery, as well as for the TsA induced upregulation of 5-LO promoter activity. TsA does not alter the protein expression levels of Sp1 and Sp3 as investigated in Western blot analysis, neither in HeLa nor in MM6 cells. DNA affinity purification assays revealed that TsA had no effect on the DNA affinity of Sp1 or Sp3. In vitro binding of both Sp1 and Sp3 to the 5-fold GC box, GC4 and GC5 was demonstrated by DAPA analysis, but histone deacetylase inhibition did not change the associated protein amounts. Finally, in vivo binding of Sp1 and Sp3 was investigated in chromatin immunoprecipitation assay (ChIP) in MM6 cells. TsA clearly induced the association of both proteins to the promoter area surrounding the TIS. Upon TsA treatment also RNA polymerase II binding to the area surrounding the TIS (-318 to +52 bp) was increased and even initiated in the more distal promoter parts –1049 to –292 bp, which are negatively regulated in reporter gene assays. Interestingly histone H4 is already highly acetylated without TsA treatment and the acetylation status of H4 remains unchanged after histone deacetylase inhibition, indicating an open chromatin structure of the 5-LO gene in MM6 cells. In a cotransfection study with Sp1 and Sp3, the transactivating potential of factors was investigated and in accordance with the ChIP data, Sp1 and Sp3 increased the promoter activity, but only after TsA treatment. In gel shift assays, the influence of DNA methylation on Sp1 binding was investigated. The results indicate different roles for the three proximal promoter sites. Whereas Sp1 binding to the 5-fold GC box and GC4 is impaired by DNA methylation, binding to GC5 is even increased. A cotransfection study with methylated 5-LO promoter constructs and the murine methyl-CpG binding proteins suggest MBD1 involvement in the regulation of the 5-LO promoter. Since in gel shifts Sp1 binding is inhibited by DNA methylation, at least to the 5-fold GC box and the activating element GC4, and similarly the mutation/deletion of the same sites strongly reduces or inhibits promoter activity, it is likely to assume, that the loss of promoter activity after in vitro methylation is in the first place due to impaired Sp1/Sp3 binding. Together the data underline the importance and complexity of Sp1/Sp3 binding to the GC rich sites in the regulation of 5-LO promoter activity in response to the histone deacetylase inhibitor TsA as well as in respect to DNA methylation.
Am Anfang dieser Arbeit wurde die CE als hervorragende Methode zur Identifizierung und Charakterisierung von Nukleinsäuren und Nukleinsäure-Analoga etabliert. Die Anwendung der kommerziell erhältlichen Gelkapillaren führte zwar zu guten Trennergebnissen, zeigte jedoch erhebliche Mängel im praktischen Umgang (Giftigkeit der Agentien), eingeschränkte Lebensdauer der Gele aufgrund von Degradations-prozessen, in der Sensitivität sowie in der Reproduzierbarkeit der erhaltenen Ergebnisse. Aus diesem Grund wurde die Methode im Bezug auf die Gelbildner und die verwendete Detektionsmethode erheblich verbessert. Die Verwendung von Cellulosederivaten als Gelbildner führte zu einer extrem robuster, sensitiven, preiswerten und ungiftigen Methode zur Trennung von komplexen Nukleinsäuregemischen. Die Möglichkeit die Trennmatrix für jeden Lauf zu erneuern, verbesserte die Reproduzierbarkeit der Methode enorm. Für verschiedene Trennprobleme kann durch einfache Veränderung der Viskosität oder der Konzentration des Gelbildners die Methode einfach und schnell optimiert werden. Die etablierte Standardmethode konnte zur Detektion und Charakterisierung unterschiedlichster DNA Fragmente herangezogen werden. So konnte eine Reinheitsprüfung für Oligonukleotide eingeführt werden, Mutationen detektiert werden oder ein weiterer Parameter zur Charakterisierung von Antisense Oligonukleotide etabliert werden. Die zusätzliche Einführung der laserinduzierten Detektion (LIF) in die Standardmethode führte zu einer enormen Steigerung der Sensitivität der Methode. Hiermit können 100-fach kleinere Konzentrationen von Nukleinsäuren detektiert werden, als mit der herkömmlichen UV-Detektion. Basierend auf diesen Optimierungen ist es gelungen die entwickelte CE Methode als ein wesentlichen Bestandteil eines hochselektiven und spezifischen zellbasierten Modells zur Detektion von apoptotischen Vorgängen zu etablieren. Dieses Modell hat, im Gegensatz zu den zahlreichen, existierenden Cytotox-Assays, den entscheidenden Vorteil, dass es auf Wirkebene detektiert und nicht nur Surrogatparameter, wie zum Beispiel Zellmembranschäden, erfasst. Trotzdem ist es einfach und schnell durchführbar. Dadurch ist das etablierte Modell zur klaren Unterscheidung von Apoptose und Nekrose in der Lage. Diese klare Unterscheidung macht ein Wirkstoff-Screening von potentiell apoptose-induzierenden Wirkstoffen einfach und schnell möglich. Wie gezeigt wurde, kann durch die Verwendung von Tumorzelllinien mit verschiedenen Prädispositionen die Unterscheidung von unspezifisch induzierter Apoptose, wie z.B. durch Serumentzug, und von spezifisch induzierter Apoptose, wie z.B. durch Etoposid bei Caspase-3 enthaltenden Zelllinien und Vitamin-D-Analoga bei Vitamin-D3-Rezeptor tragenden Tumorzelllinien, möglich gemacht werden. Das Potential des etablierten Modells kann durch Einsatz verschiedenster Tumorzellen erweitert werden. So kann man die Wirksamkeit sowie die Resistenz der Tumorzellen gegenüber vorhandenen und neuen Tumortherapeutika ohne größere Probleme zeigen.
In der vorliegenden Arbeit wurden neue Interaktionspartner des einwärtsrektifizierenden, renalen ROMK-Kaliumkanals identifiziert und funktionell in Xenopus Oozyten untersucht. Zunächst wurde mit Hilfe eines modifizierten Hefe-Zwei-Hybrid-Systems und dem zytosolischen C-Terminus von ROMK als Köderprotein eine cDNS-Bibliothek der humanen Niere durchmustert. Eine Besonderheit hierbei war, daß das Köderprotein im Gegensatz zu dem herkömmlichen Hefe-Zwei-Hybrid-System in der nativen, tetrameren Konformation vorlag. Die Interaktion der isolierten Proteine mit dem ROMK-C-Terminus wurde anschließend in der Hefe in direkten Bindungsstudien bestätigt. Auf diese Weise konnten 25 neue Interaktionspartner für ROMK gefunden werden. Aufgrund ihrer teilweise bekannten Funktionen und Strukturen wurden einige, insbesondere das Golgi-Protein Golgin-160, das Adapterprotein GRB7 und die Serin/Threonin-Proteinphosphatase-Untereinheit PP2A B56β, für eine weitergehende Charakterisierung ausgewählt. Die vermutete Beteiligung von Golgin-160 am vesikulären Membrantransport machte die gefundene Interaktion mit ROMK besonders interessant, da über den Transport des Kanals vom Endoplasmatischen Retikulum über den Golgi-Apparat bis an die Zelloberfläche nur wenig bekannt ist. Zunächst konnte die Bindung von Golgin-160 an das ROMK Kanalprotein durch Koimmunpräzipitation beider Proteine aus Lysaten transfizierter Säugerzellen unterstützt werden. Immunfluoreszenzmikroskopische Untersuchungen bestätigten weiterhin, daß beide Proteine tatsächlich und ausschließlich im Bereich des Golgi-Apparats kolokalisiert sind. Dies verstärkte die Vermutung, daß Golgin-160 am Membrantransport von ROMK beteiligt ist. Funktionelle Untersuchungen in Xenopus Oozyten mit Hilfe der Zwei-Elektroden-Spannungsklemme ergaben nach Koexpression beider Proteine reproduzierbar eine Verdopplung der ROMK-Stromamplitude. Mittels einer Chemolumineszenz-Oberflächenexpressionsanalyse konnte dies auf eine Zunahme der Dichte des Kanalproteins in der Plasmamembran zurückgeführt werden. Ähnliche Resultate wurden auch für das nahe verwandte Kir2.1-Kanalprotein erhalten. Diese Untersuchungen zeigten zudem, daß nur das Kanalprotein an der Zelloberfläche, nicht aber die Gesamtmenge des Proteins in der Zelle erhöht war. Dementsprechend waren auch die biophysikalischen und pharmakologischen Eigenschaften des ROMK-Kanals durch das koexprimierte Golgin-160 nicht verändert. Um die Bedeutung der gefundenen Interaktion näher zu untersuchen, wurden Bindungsstudien mit C-terminalen Bartter-Mutanten von ROMK durchgeführt. Alle untersuchten Punkt- und Trunkationsmutanten waren noch zur Bindung von Golgin-160 fähig, und zwei Punktmutanten konnten durch Golgin-160 auch funktionell stimuliert werden. Daraus kann geschlossen werden, daß diese hochkonservierten Aminosäurereste des Kanalproteins nicht an der Bindung von Golgin-160 beteiligt sind, und daß der defekte Membrantransport dieser krankheitsverursachenden Mutanten nicht auf einer gestörten Interaktion mit dem untersuchten Golgi-Protein beruht. Mit diesen Untersuchungen wurde erstmalig gezeigt, daß Golgin-160 am Golgi-Apparat selektiv mit transportierten Membranproteinen interagiert und dadurch deren Zelloberflächenexpression reguliert. Eine spezifische Rolle beim Transport von Oberflächenproteinen zur Plasmamembran wird durch das Ergebnis unterstrichen, daß auch die Oberflächenexpression der entfernt verwandten Kv1.5- und Kv4.3-Kanalproteine stimuliert wird, aber nicht die des HERGKaliumkanals. In weiteren funktionellen Untersuchungen konnten auch für GRB7 und PP2A B56β erstmalig Einflüsse auf die ROMK-Kanalaktivität gezeigt werden. Die Koexpression von GRB7 führte sowohl bei ROMK als auch bei verwandten Kir2-Kanalproteinen zu einer Verringerung der Stromamplitude. Bei PP2A B56β war der Effekt von der Expressionshöhe dieser regulatorischen Phosphatase- Untereinheit abhängig. So waren die ROMK-Ströme bei geringen Mengen an injizierter PP2A B56β erhöht, nach Injektion größerer Mengen dagegen reduziert. Die Regulation der ROMK-Kanalaktivität wird größtenteils durch die Kontrolle der Kanaldichte an der Zelloberfläche erzielt. Da unterschiedliche Signalwege die Häufigkeit des Kanalproteins an der Zelloberfläche modulieren können, kann vermutet werden, daß nicht nur Golgin-160 sondern auch GRB7 und PP2A B56β an der Regulation der Oberflächenexpression von ROMK beteiligt sind (Abb. 36). Die Identifizierung dieser neuen Interaktionspartner stellt deshalb einen ersten wichtigen Schritt bei der Aufklärung der dieser Regulation zugrundeliegenden molekularen Mechanismen dar.
The goal of this thesis was the development, evaluation and application of novel virtual screening approaches for the rational compilation of high quality pharmacological screening libraries. The criteria for a high quality were a high probability of the selected molecules to be active compared to randomly selected molecules and diversity in the retrieved chemotypes of the selected molecules to be prepared for the attrition of single lead structures. For the latter criterion the virtual screening approach had to perform “scaffold hopping”. The first molecular descriptor that was explicitly reported for that purpose was the topological pharmacophore CATS descriptor, representing a correlation vector (CV) of all pharmacophore points in a molecule. The representation is alignment-free and thus renders fast screening of large databases feasible. In a first series of experiments the CATS descriptor was conceptually extended to the three-dimensional pharmacophore-pair CATS3D descriptor and the molecular surface based SURFCATS descriptor. The scaling of the CATS3D descriptor, the combination of CATS3D with different similarity metrics and the dependence of the CATS3D descriptor on the threedimensional conformations of the molecules in the virtual screening database were evaluated in retrospective screening experiments. The “scaffold hopping” capabilities of CATS3D and SURFCATS were compared to CATS and the substructure fingerprint MACCS keys. Prospective virtual screening with CATS3D similarity searching was applied for the TAR RNA and the metabotropic glutamate receptor 5 (mGlur5). A combination of supervised and unsupervised neural networks trained on CATS3D descriptors was applied prospectively to compile a focused but still diverse library of mGluR5 modulators. In a second series of experiments the SQUID fuzzy pharmacophore model method was developed, that was aimed to provide a more general query for virtual screening than the CATS family descriptors. A prospective application of the fuzzy pharmacophore models was performed for TAR RNA ligands. In a last experiment a structure-/ligand-based pharmacophore model was developed for taspase1 based on a homology model of the enzyme. This model was applied prospectively for the screening for the first inhibitors of taspase1. The effect of different similarity metrics (Euc: Euclidean distance, Manh: Manhattan distance and Tani: Tanimoto similarity) and different scaling methods (unscaled, scaling1: scaling by the number of atoms, and scaling2: scaling by the added incidences of potential pharmacophore points of atom pairs) on CATS3D similarity searching was evaluated in retrospective virtual screening experiments. 12 target classes of the COBRA database of annotated ligands from recent scientific literature were used for that purpose. Scaling2, a new development for the CATS3D descriptor, was shown to perform best on average in combination with all three similarity metrics (enrichment factor ef (1%): Manh = 11.8 ± 4.3, Euc = 11.9 ± 4.6, Tani = 12.8 ± 5.1). The Tanimoto coefficient was found to perform best with the new scaling method. Using the other scaling methods the Manhattan distance performed best (ef (1%): unscaled: Manh = 9.6 ± 4.0, Euc = 8.1 ± 3.5, Tani = 8.3 ± 3.8; scaling1: Manh = 10.3 ± 4.1, Euc = 8.8 ± 3.6, Tani = 9.1 ± 3.8). Since CATS3D is independent of an alignment, the dependence of a “receptor relevant” conformation might also be weaker compared to other methods like docking. Using such methods might be a possibility to overcome problems like protein flexibility or the computational expensive calculation of many conformers. To test this hypothesis, co-crystal structures of 11 target classes served as queries for virtual screening of the COBRA database. Different numbers of conformations were calculated for the COBRA database. Using only a single conformation already resulted in a significant enrichment of isofunctional molecules on average (ef (1%) = 6.0 ± 6.5). This observation was also made for ligand classes with many rotatable bonds (e.g. HIV-protease: 19.3 ± 6.2 rotatable bonds in COBRA, ef (1%) = 12.2 ± 11.8). On average only an improvement from using the maximum number of conformations (on average 37 conformations / molecule) to using single conformations of 1.1 fold was found. It was found that using more conformations actives and inactives equally became more similar to the reference compounds according to the CATS3D representations. Applying the same parameters as before to calculate conformations for the crystal structure ligands resulted in an average Cartesian RMSD of the single conformations to the crystal structure conformations of 1.7 ± 0.7 Å. For the maximum number of conformations, the RMSD decreased to 1.0 ± 0.5 Å (1.8 fold improvement on average). To assess the virtual screening performance and the scaffold hopping potential of CATS3D and SURFACATS, these descriptors were compared to CATS and the MACCS keys, a fingerprint based on exact chemical substructures. Retrospective screening of ten classes of the COBRA database was performed. According to the average enrichment factors the MACCS keys performed best (ef (1%): MACCS = 17.4 ± 6.4, CATS = 14.6 ± 5.4, CATS3D = 13.9 ± 4.9, SURFCATS = 12.2 ± 5.5). The classes, where MACCS performed best, consisted of a lower average fraction of different scaffolds relative to the number of molecules (0.44 ± 0.13), than the classes, where CATS performed best (0.65 ± 0.13). CATS3D was the best performing method for only a single target class with an intermediate fraction of scaffolds (0.55). SURFCATS was not found to perform best for a single class. These results indicate that CATS and the CATS3D descriptors might be better suited to find novel scaffolds than the MACCS keys. All methods were also shown to complement each other by retrieving scaffolds that were not found by the other methods. A prospective evaluation of CATS3D similarity searching was done for metabotropic glutamate receptor 5 (mGluR5) allosteric modulators. Seven known antagonists of mGluR5 with sub-micromolar IC50 were used as reference ligands for virtual screening of the 20,000 most drug-like compounds – as predicted by an artificial neural network approach – of the Asinex vendor database (194,563 compounds). Eight of 29 virtual screening hits were found with a Ki below 50 µM in a binding assay. Most of the ligands were only moderately specific for mGluR5 (maximum of > 4.2 fold selectivity) relative to mGluR1, the most similar receptor to mGluR5. One ligand exhibited even a better Ki for mGluR1 than for mGluR5 (mGluR5: Ki > 100 µM, mGluR1: Ki = 14 µM). All hits had different scaffolds than the reference molecules. It was demonstrated that the compiled library contained molecules that were different from the reference structures – as estimated by MACCS substructure fingerprints – but were still considered isofunctional by both CATS and CATS3D pharmacophore approaches. Artificial neural networks (ANN) provide an alternative to similarity searching in virtual screening, with the advantage that they incorporate knowledge from a learning procedure. A combination of artificial neural networks for the compilation of a focused but still structurally diverse screening library was employed prospectively for mGluR5. Ensembles of neural networks were trained on CATS3D representations of the training data for the prediction of “mGluR5-likeness” and for “mGluR5/mGluR1 selectivity”, the most similar receptor to mGluR5, yielding Matthews cc between 0.88 and 0.92 as well as 0.88 and 0.91 respectively. The best 8,403 hits (the focused library: the intersection of the best hits from both prediction tasks) from virtually ranking the Enamine vendor database (ca. 1,000,000 molecules), were further analyzed by two self-organizing maps (SOMs), trained on CATS3D descriptors and on MACCS substructure fingerprints. A diverse and representative subset of the hits was obtained by selecting the most similar molecules to each SOM neuron. Binding studies of the selected compounds (16 molecules from each map) gave that three of the molecules from the CATS3D SOM and two of the molecules from the MACCS SOM showed mGluR5 binding. The best hit with a Ki of 21 µM was found in the CATS3D SOM. The selectivity of the compounds for mGluR5 over mGluR1 was low. Since the binding pockets in the two receptors are similar the general CATS3D representation might not have been appropriate for the prediction of selectivity. In both SOMs new active molecules were found in neurons that did not contain molecules from the training set, i. e. the approach was able to enter new areas of chemical space with respect to mGluR5. The combination of supervised and unsupervised neural networks and CATS3D seemed to be suited for the retrieval of dissimilar molecules with the same class of biological activity, rather than for the optimization of molecules with respect to activity or selectivity. A new virtual screening approach was developed with the SQUID (Sophisticated Quantification of Interaction Distributions) fuzzy pharmacophore method. In SQUID pairs of Gaussian probability densities are used for the construction of a CV descriptor. The Gaussians represent clusters of atoms comprising the same pharmacophoric feature within an alignment of several active reference molecules. The fuzzy representation of the molecules should enhance the performance in scaffold hopping. Pharmacophore models with different degrees of fuzziness (resolution) can be defined which might be an appropriate means to compensate for ligand and receptor flexibility. For virtual screening the 3D distribution of Gaussian densities is transformed into a two-point correlation vector representation which describes the probability density for the presence of atom-pairs, comprising defined pharmacophoric features. The fuzzy pharmacophore CV was used to rank CATS3D representations of molecules. The approach was validated by retrospective screening for cyclooxygenase 2 (COX-2) and thrombin ligands. A variety of models with different degrees of fuzziness were calculated and tested for both classes of molecules. Best performance was obtained with pharmacophore models reflecting an intermediate degree of fuzziness. Appropriately weighted fuzzy pharmacophore models performed better in retrospective screening than CATS3D similarity searching using single query molecules, for both COX-2 and thrombin (ef (1%): COX-2: SQUID = 39.2., best CATS3D result = 26.6; Thrombin: SQUID = 18.0, best CATS3D result = 16.7). The new pharmacophore method was shown to complement MOE pharmacophore models. SQUID fuzzy pharmacophore and CATS3D virtual screening were applied prospectively to retrieve novel scaffolds of RNA binding molecules, inhibiting the Tat-TAR interaction. A pharmacophore model was built up from one ligand (acetylpromazine, IC50 = 500 µM) and a fragment of another known ligand (CGP40336A), which was assumed to bind with a comparable binding mode as acetylpromazine. The fragment was flexible aligned to the TAR bound NMR conformation of acetylpromazine. Using an optimized SQUID pharmacophore model the 20,000 most druglike molecules from the SPECS database (229,658 compounds) were screened for Tat-TAR ligands. Both reference inhibitors were also applied for CATS3D similarity searching. A set of 19 molecules from the SQUID and CATS3D results was selected for experimental testing. In a fluorescence resonance energy transfer (FRET) assay the best SQUID hit showed an IC50 value of 46 µM, which represents an approximately tenfold improvement over the reference acetylpromazine. The best hit from CATS3D similarity searching showed an IC50 comparable to acetylpromazine (IC50 = 500 µM). Both hits contained different molecular scaffolds than the reference molecules. Structure-based pharmacophores provide an alternative to ligand-based approaches, with the advantage that no ligands have to be known in advance and no topological bias is introduced. The latter is e.g. favorable for hopping from peptide-like substrates to drug-like molecules. A homology model of the threonine aspartase taspase1 was calculated based on the crystal structures of a homologous isoaspartyl peptidase. Docking studies of the substrate with GOLD identified a binding mode where the cleaved bond was situated directly above the reactive N-terminal threonine. The predicted enzyme-substrate complex was used to derive a pharmacophore model for virtual screening for novel taspase1 inhibitors. 85 molecules were identified from virtual screening with the pharmacophore model as potential taspase1- inhibitors, however biochemical data was not available before the end of this thesis. In summary this thesis demonstrated the successful development, improvement and application of pharmacophore-based virtual screening methods for the compilation of molecule-libraries for early phase drug development. The highest potential of such methods seemed to be in scaffold hopping, the non-trivial task of finding different molecules with the same biological activity.
Boswellia serrata gum resin extracts (frankincense) have been used for centuries in folk medicine in Asia and Africa. They have shown beneficial therapeutic effects, particularly in the treatment of chronic inflammatory diseases. Clinical studies on humans confirmed an anti-inflammatory and anti-cancer potential of Frankincense preparations. Boswellic acids (BAs) are the major ingredients, responsible for the pharmacological action of the extracts. Molecular and cellular studies with BAs revealed a number of targets including 5-lipoxygenase (LO), topoisomerases and the NF-κB pathway. Since there is little information on the modulation of cellular physiology by BAs, this work was designed to provide a detailed investigation of the cellular and molecular effects of BAs in several cell types related to inflammation. We report that 11-keto-BAs are potent activators of functional responses in human neutrophils, a type of leukocytes mediating acute inflammatory processes. Neutrophil activation by 11-keto-BAs is reflected by enhanced generation of oxygen radicals, release of arachidonic acid (AA) and the subsequent transformation of AA to pro-inflammatory eicosanoids. Investigation of the participating signalling pathways identified Ca2+, phosphoinositide-3 kinase, and members of the MAP kinase family (ERKs) as mediators. Second, we present a detailed study of the modulation of human platelet physiology and intracellular signalling events by BAs. Intriguingly, we discovered an inverse structure-activity relationship of BAs regarding platelet activation, with 11-methylene-BAs being superior over 11-keto-BAs. Thus, 11-methylene-BAs stimulated platelet Ca2+ mobilisation, MAP kinase and Akt activation, AA release, 12-LO and cyclooxygenase product formation, and thrombin generation. Novel Ca2+-independent activation pathways of platelet lipid metabolism were discovered. In contrast, 11-keto-BAs were inactive but found to inhibit platelet (p)12-LO directly. Interaction with p12-LO was confirmed in a pulldown assay using immobilised BAs as bait. Finally, BAs were shown to attenuate the activation of monocytes, a cell type responsible for the maintenance of chronic inflammatory states. Impairment of Ca2+ homeostasis is likely conferred by inhibition of Ca2+ influx channels. Taken together, our results shed light on the modulation of intracellular physiology of inflammatory cells by BAs, contributing to a better understanding of the anti-inflammatory effects exerted by frankincense preparations.
Alzheimer's disease-related mutations in the presenilin-1 gene (PS1) are leading to an elevated production of neurotoxic beta-amyloid 1-42 and may additionally enhance oxidative stress. Here, we provide in vivo evidence indicating that brains of transgenic mice expressing different human Alzheimer-linked PS1 mutations exhibit a reduced activity of two antioxidant enzymes. For this purpose, mice transgenic for human PS1 and for single and multiple PS1 mutations were generated. Mice with multiple PS1 mutations showed a significantly decreased activity of the antioxidant enzymes Cu/Zn superoxide dismutase and glutathione reductase already at an age of 3-4 months. As expected, this effect was less pronounced for the mice with a single PS1 mutation. By contrast, animals bearing normal human PS1 showed significantly elevated enzyme activities relative to non-transgenic littermate controls.
Enhanced apoptosis and elevated levels of reactive oxygen species (ROS) play a major role in aging. In addition, several neurodegenerative diseases are associated with increased oxidative stress and apoptosis in neuronal tissue. Antioxidative treatment has neuro-protective effects. The aim of the present study was to evaluate changes of susceptibility to apoptotic cell death by oxidative stress in aging and its inhibition by the antioxidant Ginkgo biloba extract EGb761. We investigated basal and ROS-induced levels of apoptotic lymphocytes derived from the spleen in young (3 months) and old (24 months) mice. ROS were induced by 2-deoxy-D-ribose (dRib) that depletes the intracellular pool of reduced glutathione. Lymphocytes from aged mice accumulate apoptotic cells to a significantly higher extent under basal conditions compared to cells from young mice. Treatment with dRib enhanced this difference, implicating a higher sensitivity to ROS in aging. Apoptosis can be reduced in vitro by treatment with EGb761. In addition, mice were treated daily with 100mg/kg EGb761 per os over a period of two weeks. ROS-induced apoptosis was significantly reduced in the EGb761 group. Interestingly, this effect seemed to be more pronounced in old mice.
Apoptosis seems to be involved in immunosenescence associated with aging. Moreover, in lymphocytes (PBL) of patients with Alzheimer's disease, an increased susceptibility to the apoptotic pathway has been described possibly due to impaired protection of oxidative stress. Accordingly, it seemed to be of particular interest to investigate the contribution of normal aging to the susceptibility from human lymphocytes to programmed cell death. We could show that PBL from elderly individuals (>60 years) accumulate apoptosing cells to a significant higher extent in spontaneous and activation-induced cell death compared to younger controls (<35 years). Treatment with the oxidative stressor 2-deoxy-D-ribose or with agonistic-CD95-antibody pronounced this effect even more implicating a higher sensitivity to reactive oxygen species and a higher functional CD95 expression, respectively. In addition, expression of the activation markers HLA-DR and CD95 was significantly increased in CD3+-cells of aged subjects, while expression of CD25 did not seem to be affected by age. Expression of Bcl-2 was increased in aging and correlated with the number of apoptotic cells.
The relevance of physiological immune aging is of great interest with respect to determining disorders with pathologic immune function in aging individuals. In recent years, the relevance of changes in peripheral lymphocytes in age-associated neurologic diseases has become more evident. Due to the lack of immunological studies, covering more than one event after mitogenic activation, we envisaged a new concept in the present study, aiming to investigate several events, starting from T cell receptor (TCR) ligation up to T cell proliferation. In addition, we addressed the question whether changes are present in the subsets (CD4, CD8) with aging. Phosphorylation of tyrosine residues declines with increasing age in CD4+ cells. Fewer levels of CD69 positive cells after 4 h mitogenic activation, altered expression of cytokines (IL2, IFN-gamma and TNF-alpha; 22 h) and lower proliferation (72 h) were determined in aging. Moreover, it could be shown that CD8+ lymphocytes react more effectively to mitogenic stimulation with reference to CD69 expression and proliferation in both age groups (<35 and >60 years old). These data indicate that T cell activation, mediated by TCR engagement, is significantly impaired in aging and both subsets are affected. However, bypassing the TCR does not fully restore T cell function, indicating that there are more mechanisms involved than impaired signal transduction through TCR only. The results will be discussed in relation to their relevance in neurodegenerative and psychiatric disorders.
In vivo manipulation of interleukin-2 expression by a retroviral tetracycline (tet)-regulated system

We have used the tetracycline (tet)-regulated system as described previously to evaluate the applicability of controlled gene expression in cancer gene therapy. As a model gene, we used the human interleukin-2 (IL-2) gene, which has been placed under the transcriptional control of the tetO/promoter. Human melanoma cells were transduced by two modified retroviral tet vectors containing the transactivator regulatory unit and the IL-2 gene driven by the tetO/promoter, respectively. In the absence of tet, IL-2 expression in the target cells was stable over several months. IL-2 production was in the range of 40 U/106 cells/24 hours. A fine tuning of IL-2 expression could be achieved by culturing the transduced cells with increasing doses of tet, whereby a concentration of 500 ng/mL tet in the culture medium abrogated IL-2 expression. Most importantly for clinical application, IL-2 expression by the transduced melanoma cells could also be regulated in vivo. When nu/nu mice were inoculated with the transduced tumor cells, they failed to develop tumors. Instead, the inhibition of IL-2 expression in the transduced tumor cells by oral administration of tet led to subcutaneous tumor growth; this growth rate was comparable with the growth rate of subcutaneously inoculated untransduced parental cells. The finding demonstrates the applicability of the tet-regulated system in cancer gene therapy.
Many cases of early-onset inherited Alzheimer's disease (AD) are caused by mutations in the presenilin-1 (PS1) gene. Expression of PS1 mutations in cell culture systems and in primary neurons from transgenic mice increases their vulnerability to cell death. Interestingly, enhanced vulnerability to cell death has also been demonstrated for peripheral lymphocytes from AD patients. We now report that lymphocytes from PS1 mutant transgenic mice show a similar hypersensitivity to cell death as do peripheral cells from AD patients and several cell culture systems expressing PS1 mutations. The cell death-enhancing action of mutant PS1 was associated with increased production of reactive oxygen species and altered calcium regulation, but not with changes of mitochondrial cytochrome c. Our study further emphasizes the pathogenic role of mutant PS1 and may provide the fundamental basis for new efforts to close the gap between studies using neuronal cell lines transfected with mutant PS1, neurons from transgenic animals, and peripheral cells from AD patients. Methoden zur Präparation und Charakterisierung von time dependent haze auf Siliziumoberflächen

Zwischen der Herstellung von Siliziumwafern und ihrer weiteren Verarbeitung zu Halbleiterbauelementen liegt eine Zeitspanne, in welcher die Scheiben in Kunststoffboxen aufbewahrt werden. Während dieser Lagerung kann es zu einem Ansteigen der Oberflächenrauhigkeit kommen. Dieses Phänomen wird als Time Dependent Haze (TDH) bezeichnet und durch chemische Prozesse von Verunreinigungen auf der Siliziumoberfläche verursacht. Diese Kontaminationen entstammen dem Kunststoff der Transportboxen, der Reinraumluft oder den Chemikalien der naßchemischen Waferreinigung. Time Dependent Haze wurde künstlich hergestellt und charakterisiert. Die Präparationen erfolgten in verschiedenen Versuchsreihen mit organischen und anorganischen Verbindungen. Diese gezielten Kontaminationen erlauben Rückschlüsse auf Substanzen und Einflußgrößen, welche TDH verursachen. Nach einer Lagerungsperiode wurden die Siliziumoberflächen mittels Streulichtmessungen untersucht. Das Auftreten von Licht-Punkt-Defekten diente als Maßstab für die TDH-Bildung. Mit verschiedenen analytischen Methoden, insbesondere dem Rasterkraftmikroskop, wurde TDH in einem frühen Entstehungszustand untersucht. Präparationen mit flüchtigen organischen Verbindungen zeigten, daß solche Substanzen TDH erzeugten, die sowohl einen hohen Dampfdruck als auch eine gute Wasserlöslichkeit besitzen. Beide Eigenschaften sind notwendig, um über die Gasphase auf die hydrophile Oberfläche eines Wafers zu gelangen. Die Morphologie dieses TDH fiel sehr unterschiedlich aus. Im Fall der Präparation mit Aceton enstanden Partikel von wenigen Nanometern Durchmesser. Die gezielte Verunreinigung mit Tetrahydrofuran führte zu einem Kontaminationsfilm mit einer sehr geringen Mikrorauhigkeit. Anorganische Verbindungen wie beispielsweise Schwefelsäure, die in der Waferreinigung Verwendung findet, können bei fehlerhaften Reinigungsschritten TDH bilden. Unter dem Einfluß einer solchen Lösung, bestehend aus H2SO4/H2O2, entstanden sulfathaltige Kristallite, welche im Verlauf der Waferlagerung agglomerierten. Das gleiche Resultat ergab eine Präparation, bei der Siliziumwafer einer Atmosphäre aus Schwefeldioxid ausgesetzt waren. Sehr wahrscheinlich existieren die Kristallisationskeime bereits unmittelbar nach der Präparation. Aufgrund ihrer geringen Größe können diese von der Streulichtmessung nicht als Partikel erfaßt werden. Erst nach mehr monatiger Waferlagerung ist das Kristallwachstum soweit fortgeschritten, daß das untere Detektionslimit der Streulichtmethode erreicht wird und die Partikel als solche registriert werden können. Auf Wafern mit organischem TDH konnte Kupfer nachgewiesen werden. Dieses Metall ist als Verunreinigung im Reinstsilizium enthalten. Es diffundiert innerhalb der Lagerungsperiode zur Oberfläche und scheidet sich ab. Dort fungiert Kupfer als Nukleationszentrum für Kontaminanten und fördert auf diese Weise die TDH-Entstehung. Experimente mit mehreren Einflußgrößen, bei welchen Siliziumwafer verschiedenen Parametern gleichzeitig ausgesetzt waren, zeigten synergetische Effekte in Bezug auf die Bildung von Time Dependent Haze. Im Vergleich zu Präparationen mit nur einem Kontaminanten lag die Zahl der neu entstandenen Licht-Punkt-Defekte unerwartet hoch. Innerhalb dieser Versuchsmatrix hatten die Anwesenheit von Ammoniumsulfat und eine hohe Luftfeuchtigkeit einen deutlichen Einfluß auf die TDH-Entstehung. Die Siliziumoberfläche wird durch einzelne Substanzen aktiviert und die Abscheidung weiterer Kontaminationen dadurch erhöht. Eine nähere Untersuchung von TDH bestehend aus verschiedenen Kontaminanten erfolgte mit Hilfe des Pulsed Force Mode/AFM. Diese Methode ermöglicht anhand der Bestimmung der Adhäsion zwischen einer AFM-Meßspitze und der Oberfläche die Unterscheidung verschiedener Materialien mit der hohen Auflösung eines Rasterkraftmikroskops. Zu Beginn der Untersuchungen erfolgte eine Evaluation der Methode. Dabei zeigte sich, daß die Adhäsion unbeeinflußt von der Partikelgröße ist. Das Adhäsionssignal ist abhängig vom Radius der AFM-Spitze und von der umgebenden Luftfeuchtigkeit. Dieses Signal setzt sich zusammen aus der Oberflächenenergie des Materials und aus dem Wasser- und Kontaminationsfilm, welcher die Oberfläche bedeckt. Adhäsionsmessungen an kontaminierten Wafern zeigten außer der rasterkraftmikroskopischen Abbildung der Oberfläche zusätzliche Strukturen. Diese deuten auf das gleichzeitige Auftreten von TDH-Partikeln mit unterschiedlicher Zusammensetzung hin. Die Ergebnisse dieser Arbeit bestätigten ein bereits existierendes Modell zur Entstehung von Time Dependent Haze. Unter Berücksichtigung der neuen Erkenntnisse konnte dieses Modell erweitert werden.
Im Rahmen dieser Arbeit sollte ein Anti-PGE2-Antikörperfragment exprimiert werden, mit dem über NMR-Spektroskopie genauere Strukturdaten der Antikörperbindungsdomäne zu erhalten wären. Da Fab-, Fv- und scFv-Fragmente die Antigenbindungsstelle besitzen und sich gegenüber einem vollständigem IgG zusätzlich durch ihre geringere Größe auszeichnen, wurden die korrespondierenden Antikörperfragmente für diese Arbeiten gewählt. Ausgehend von einer etablierten Hybridom-Zelllinie konnten über cDNA-Synthese und Assembly-PCR die Gene für die Antikörperfragmente bereitgestellt werden. Zum Verifizieren von DNA-Sequenz und Bindungseigenschaften stand ein Anti-PGE2-Fab-Fragment früherer Arbeiten von Frau Dr. Ilse Zündorf zur Verfügung. Für Strukturuntersuchungen müssen relativ große Proteinmengen bereit gestellt werden, daher ist es notwendig, hohe Ausbeuteverluste durch Präzipitation, Degradierung, Aufreinigungsverluste u.a. zu vermeiden. Um zunächst eine möglichst gute Proteinexpression zu erhalten, wurde der Bakterienstamm Escherichia coli BL21DE3 und das pET-Expressionssystem gewählt. In diesem System war zu erwarten, dass die überexprimierten Antikörperfragmente unter den gewählten Bedingungen in Form unlöslicher Aggregate, sogenannter inclusion bodies, in den Zellen vorliegen werden. Die der Klonierung der VH-, Vk- und scFv-Gene folgende Expression der Proteine zeigte, dass es tatsächlich zur Bildung von inclusion bodies kommt. Die Vorteile dieser Aggregatbildung, wie hohe Proteinmenge, Proteaseunempfindlichkeit aufgrund der hohen Dichte sowie die hohe Homogenität, sollten für die Gewinnung nativer Antikörperfragmente genutzt werden. Hierfür konnte ein System etabliert werden, das im wesentlichen auf der Solubilisierung der Aggregate und der Reduktion falscher Disulfidbrücken mit anschließender Rückfaltung und Rekonstituierung der Disulfidbrücken beruht. Kritische Parameter, die den Rückfaltungsprozess beeinflussen, wurden untersucht. Hierunter fallen insbesondere die Zusammensetzung des Renaturierungssystems wie Pufferbedingungen, das Verhältnis von oxidierten zu reduzierten Glutathion und der Zusatz von sogenannten Faltungsenhancern, die Additiva. Für fast alle wesentlichen Punkte konnten Systemkomponenten etabliert werden, die für das scFv-Fragment eine nahezu vollständige Überführung der inclusion bodies in die lösliche Form erlauben. Auch eine Bindung an das Antigen Prostaglandin E2 konnte unter Anwendung von Radioimmunoassays gezeigt werden. Problematisch ist die Aufkonzentrierung der in niedriger Konzentration vorliegenden renaturierten scFv-Fragmente. Je stärker aufkonzentriert wird, desto stärker präzipitieren die scFv-Moleküle in der Ultrafiltrations- bzw. Zentrifugationsfiltereinheit. Antikörper und Antikörperframente sind ohne Frage eine sehr wichtige Molekülgruppe von molekularbiologischen und biochemischen Interesse, nicht nur in dieser Arbeitsgruppe. Im Rahmen einer möglichen Weiterführung dieser Arbeit müsste geklärt werden, ob das Präzipitationsverhalten der scFv-Fragmente eine inhärente Eigenschaft des Anti-PGE2 scFv ist, oder ob hier doch eine zumindest teilweise inkorrekte Faltung der Proteinkette vorliegt. Mögliche weitere Experimente könnten in der Variation des Linkers liegen oder das etablierte Verfahren an einem weiteren scFv-Fragment zu testen. Da die genannten Probleme bei der Aufkonzentrierung der Proteinlösung auftreten, sollten statt der angewandten Techniken alternative Methoden geprüft werden. Auch wäre es interessant zu sehen, ob sich ein z.B. kommerziell verfügbares scFv-Fragment, dessen Bindungseigenschaften bekannt sind, nach Denaturierung wieder in die native Form überführen lässt. Abschließend lässt sich sagen, dass die Herstellung von nativen Protein aus inclusion bodies viele Vorteile bietet, die für die Präparation größerer Proteinmengen nicht ungenutzt bleiben sollten.
In large models of neuronal cell death, there is a tight correlation between Cdk5 deregulation and cell-cycle dysfunction. However, pathways that link Cdk5 to the cell cycle during neuronal death are still unclear. We have investigated the molecular events that precede p25/Cdk5-triggered neuronal death using a neuronal cell line that allows inducible p25 expression. In this system, no sign of apoptosis was seen before 24 hours of p25 induction. Thus, at that time, cell-cycle-regulatory proteins were analysed by immunoblotting and some of them showed a significant deregulation. Interestingly, after time-course experiments, the earliest feature correlated with p25 expression was the phosphorylation of the retinoblastoma protein (Rb). Indeed, this phosphorylation was observed 6 hours after p25 induction and was abolished in the presence of a Cdk5 inhibitor, roscovitine, which does not inhibit the usual Rb cyclin-D kinases Cdk4 and Cdk6. Furthermore, analyses of levels and subcellular localization of Cdk-related cyclins did not reveal any change following Cdk5 activation, arguing for a direct effect of Cdk5 activity on Rb protein. This latter result was clearly demonstrated by in vitro kinase assays showing that the p25-Cdk5 complex in our cell system phosphorylates Rb directly without the need for any intermediary kinase activity. Hence, Rb might be an appropriate candidate that connects Cdk5 to cell-cycle deregulation during neuronal cell death.
The identification of specific genetic (presenilin-1 [PS1] and amyloid precursor protein [APP] mutations) and environmental factors responsible for Alzheimer's disease (AD) has revealed evidence for a shared pathway of neuronal death. Moreover, AD-specific cell defects may be observed in many other nonneuronal cells (e.g., lymphocytes). Thus, lymphocytes may serve as a cellular system in which to study risk factors of sporadic, as well as genetic AD in vivo. The aim of our present study was to clarify whether lymphocytes bearing genetic or sporadic risk factors of AD share an increased susceptibility to cell death. Additionally we examined whether a cell typespecific vulnerability pattern was present and how normal aging, the main risk factor of sporadic AD, contributes to changes in susceptibility to cell death. Here, we report that lymphocytes affected by sporadic or genetic APP and PS1 AD risk factors share an increased vulnerability to cell death and exhibit a similar cell type-specific pattern, given that enhanced vulnerability was most strongly developed in the CD4+ T-cell subtype. In this paradigm, sporadic risk factors revealed the highest impact on cell type-specific sensitivity of CD4+ T cells to apoptosis. In contrast, normal aging results in an increased susceptibility to apoptosis of both, CD4+ and CD8+ T cells.
