Retroviral vectors are powerful tools in clinical gene therapy as they integrate permanently into the target cell genome and thus guarantee long-term expression of transgenes. Therefore, they belong to the most frequently used application platforms in clinical gene therapy involving a broad range of different target cells and tissues. However, stable genomic integration of retroviral vectors can be oncogenic, as reported in several animal models and in clinical trials. In particular, γ-retroviral vectors, which derive from naturally mutagenic γ-retroviruses, integrate semirandomly into the host genome with regard to the target sequence, but have a preference for regions of active transcription and regulatory elements of transcriptionally active genes. The integration can result in overexpression of adjacent genes or disruption of ‘target’ gene expression. Moreover, γ-retroviral integration can cause modified transcripts and proteins through alternative or aberrant splicing or through premature termination of transcription.
Initially, the event of insertional mutagenesis and subsequent induction of leukemia by the genotoxicity of a γ-retroviral vector was described in a mouse model after genetic modification of hematopoietic stem cells (HSCs). Vector-related activation and overexpression of the oncogene ecotropic viral integration site-1 (Evi1) fostered clonal outgrowth and leukemogenesis. Additional genotoxic events of γ-retroviral vectors were observed in clinical HSC gene therapy trials for X-linked severe combined immune deficiency (SCID-X1), chronic granulomatous disease (X-CGD), and Wiskott-Aldrich Syndrome (WAS). But, genotoxicity induced by γ-retroviral vectors has never been described in clinical gene therapy trials involving adoptive transfer of genetically modified mature T lymphocytes. This fact is surprising, since T cells are long-lived and have a high capacity of self-renewal.
In a previous study, the susceptibility towards oncogenic transformation of mature T cells and HSCs after genetic modification was compared. It could be demonstrated that T-cell receptor (TCR)-polyclonal mature T cells are far less prone to transformation after γ-retroviral transfer of (proto-)oncogenes in vivo than HSCs. Additional experiments revealed that TCR-oligoclonal (OT-I and P14) mature T cells are transformable in the same setting and give rise to mature T-cell lymphomas (MTCLs).
In the present thesis, the susceptibility of mature T cells towards insertional mutagenesis was investigated. Within the first part of the thesis, retroviral integration sites (RISs) from 33 murine MTCLs were retrieved and subsequently analyzed in terms of integration pattern, detection of common integration sites (CIS) and gene ontology (GO). As these bioinformatic results demonstrated that insertional mutagenesis most likely contributed to mature T-cell lymphomagenesis, the susceptibility of mature T cells was directly assessed in a mouse model. Therefore, murine TCR-oligoclonal OT-I T cells were transduced with an enhanced green fluorescent protein (EGFP) encoding γ-retroviral vector and gene-modified T cells were transplanted into RAG1-/- mice. After 16 months, including one round of serial transplantation, a case of MTCL emerged. Tumor cells were characterized by CD3, CD8, TCR and ICOS expression. Integration site analysis via ligation-mediated polymerase chain reaction (LM-PCR) revealed a proviral insertion in the Janus kinase 1 (Jak1) gene. Subsequent overexpression of Jak1 could be demonstrated on transcriptional and protein level. Furthermore, T-cell lymphoma cells were characterized by an activated Jak/STAT-pathway as signal transducer and activator of transcription 3 (STAT3) was highly phosphorylated. The overexpression of Jak1 was causally implicated in tumor growth promotion as specific pharmacological inhibition of Jak1 using Ruxolitinib significantly prolonged survival of mice transplanted with these Jak1-activated tumor cells. A concluding systematic metaanalysis of available gene expression data on human mature T-cell lymphomas/leukemias confirmed the relevance of Jak/STAT overexpression in sporadic human T-cell tumorigenesis.
This was the first reported case of an insertional mutagenesis event in mature T cells in vivo. Thus, the results obtained in this thesis underline the importance of long-term monitoring of genetically modified T cells in vivo and the evaluation of vector toxicology and safety in T-cell based gene therapies. In particular, the transduction of T cells with a recombinant TCR or CAR (chimeric antigen receptor) bears a risk enhancement, as normal T-cell homeostasis is perturbed besides the general risk of insertional mutagenesis.
In this thesis the integral membrane protein diacylglycerol kinase (DAGK) from E.coli is investigated with solid-state NMR. The aim is to gain an insight into the enzyme’s mechanism through integration of kinetic, structural and dynamic data. The biological function of DAGK is the transfer of the γ-phosphate group from Mg*ATP to diacylglycerol (DAG) building phosphatidic acid (PA)[6] as port of the membrane-derived oligosaccharide cycle[31,34]. Surprisingly, DAGK does not share structural or sequential similarities with other kinases[12]. Typical sequence motives found in other kinases, which catalyze phosphoryl transfer reactions, are not found[13]. In its physiological form DAGK is a homo-trimer with nine transmembrane helices, three catalytic centers and a size of 39.6 kDa.
First, the set-up of a real-time 31P MAS NMR experiment is shown. This experiment allows measuring in real-time the simultaneous ATP hydrolysis in the aqueous phase and lipid substrate phos-phorylation in the membrane phase with atomic resolution under magic angle spinning[56]. After fast transfer of the sample into the NMR spectrometer the enzymatic reaction is started with a temperature jump. This approach of real-time MAS NMR in a dual-phase system was demonstrated for the lipid substrate analogs dioleoyl- (DOG) and dibutyrylglycerol (DBG), with a C8 and C4 aliphatic chain, respectively. The combination of 31P direct and cross polarization functions as a dynamic filter. In the 31P direct polarized experiment nuclei in both phases are detected, while in the 31P cross polar-ized experiment, only nuclei in the membrane phase are detected. Rates for substrate turnover, i.e. degradation of γP-, βP, αP-ATP and build-up of βP-, αP-ADP, free phosphate as side reaction, and PA are obtained, which reveal a Michaelis-Menten behavior with regard to Mg*ATP and DBG. Here Mg*ATP and DBG follow a random-equilibrium model, where every substrate can bind indepen-dently from the other substrate. Analyses of the peak integrals from educts and products of the enzymatic reaction, revealed the stoichiometry of the reaction: 1.5 ATP molecules are used to phos-phorylate one DBG molecule. The excess of ATP is attributed to the basal ATPase activity. Further-more, experiments with ATPγS, usually regarded as a non-hydrolysable ATP-analog, where carried out. Surprisingly, DAGK hydrolyzes ATPγS and also transfers the thio-phosphate group to the lipid acceptor DBG, which points to a certain degree of plasticity in the active center. A phosphorylated enzyme intermediate was not detected. These results suggest the building of a ternary complex of Mg*ATP, DBG and DAGK performing a direct-phosphoryl transfer reaction, without passing through a phosphorylated enzyme intermediate. Experiments with the transition state analog ortho-vanadate (Vi) showed a decoupling of the ATP hydrolysis activity from lipid substrate phosphorylation. This indicates a specific transfer site for the γ-phosphate group from ATP to DAG, which can be blocked by Vi.
A general disadvantage of NMR spectroscopy compared to other spectroscopic methods is its inherent low sensitivity. One possible starting point for the improvement of signal-to-noise per unit time is the reduction of the spin-lattice relaxation time of protons[209]. Usually 95 % of the experi-mental time is required for the relaxation of the 1H to equilibrium. The addition of paramagnetic species can be used to reduce the 1H T1[233]. In a comprehensive study four different paramagnetic agents were tested: Cu2+-EDTA, Cu2+-EDTA-tag, Gd3+-TTAHA and Gd3+-DOTA. The titration of these paramagnetic complexes showed the principle feasibility of this approach, but differences between the tested species exist. The most promising complex is Gd3+-DOTA which, at a concentration of 2 mM, causes a 10-time improvement of signal-to-noise ratio per unit time. This allowed measuring 2D 13C-13C correlation spectra of proteoliposomes in one tenth of the usual required experimental time (i.e. 10 hours vs. 4 days) with good signal-to-noise.
For the investigation of structural or dynamic changes in the protein upon substrate interaction with MAS NMR, the spectral properties CP efficiency and resolution of the DAGK in liposomes needed to be improved. The most critical step during sample preparation is the reconstitution of the membrane protein from detergent micelles into a membrane of synthetic lipids under detergent removal. For this procedure the important criteria are enzymatic activity, measured in a coupled ATPase assay[55], and homogeneity of the proteoliposomes, which was tested e.g. on a discontinuous sucrose step gradient. Therefore an extensive study was carried out, in which different detergents, lipids and lipid mixtures, techniques for detergent removal and different protein-to-lipid ratios were tested. A direct correlation between high ATPase activity and good resolution was not found. Moreover, active DAGK in a mixture of DMPC and cholesterol, which emulates the membrane features of a membrane containing DAG, showed the best CP efficiency and resolution.
The assignment of the protein backbone and amino acid side chains the first mandatory step towards the investigation of structural and dynamical features influencing and defining the enzymatic mechanism by MAS NMR. As the assignment procedure is very time consuming for a total protein, a special labeling scheme for DAGK was developed, which allows assigning most of the protein areas presumably involved in enzyme catalysis. The assignment of DAGK with solution NMR[132] was not transferable to the MAS NMR spectra. Most important for the assignment process were the unique pairs[335], two consecutive amino acids which only appear once in the amino acid sequence. These unique pairs served as anchor points. Five different multinuclear MAS NMR experiments (DARR, NCO, NCA, NCACX, NCOCX) were required for the sequential assignment. It was possible to assign 35 % of the total amino acid sequence with one sample and 8 experiments acquired at 850 MHz. The secondary structure analysis showed subtle differences to the DAGK assignment with solution NMR[132], which can be attributed to the different environment in lipid bilayers and detergent micelles.
Data about structural and dynamical changes under substrate interaction can reveal details about the enzymatic mechanism. Therefore changes in chemical shift in 2D heteronuclear correlation experiments in the apo-state and under substrate saturated conditions with the substrates Mg*AMP-PNP, a non-hydrolysable ATP-analog, DOG, a mixture of Mg*AMP-PNP and DOG as well as inhibited by Vi were recorded. The most significant peak changes were observed at the interface membrane-cytoplasm as well as the the N-terminal amphipathic helix. The residues revealing chemical shift perturbations correlate with conserved residues or such residues, for which importance for catalysis and/or folding could be shown in mutation studies[8]. Especially noticeable were the changes at the amino acids Asn 72, Lys 64, His 87, Tyr 86 and Asp 95.
Beside changes of the chemical shift, changes of line width or signal doubling were observable. These changes can point to a correlation with dynamic reorientations in the μs-ms time regime, which are most relevant for enzymatic processes. The protein backbone dynamics in the apo-state as well as saturated with the substrates or inhibited with Vi were investigated with a 15N-CODEX experiment, which is based on the reorientation of the CSA tensor upon dynamical changes[350]. Specific effects of the different substrates or analogs on the protein backbone dynamic were revealed complementing the structural data and the chemical shift perturbation experiments.
Clathrin-mediated endocytosis (CME) involves spatially and temporally restricted molecular dynamics.
Although protein kinases and the actin cytoskeleton contribute to the process, whether and how
functions of kinases and actin are integrated remains unknown. Here, we demonstrate that neural
Wiskott-Aldrich syndrome protein (N-WASP) and protein kinase CK2 form a complex and localize on
clathrin-coated vesicles (CCVs). N-WASP binds to and is phosphorylated by CK2, thereby reducing the
kinase activity of CK2. By contrast, N-WASP-promoted actin polymerization is decreased upon both
phosphorylation and binding of CK2. Knockdown of N-WASP and CK2, alone or in combination, results
in impaired endocytosis of epidermal growth factor (EGF) and increased cell-surface levels of EGF
receptor (EGFR). In order to rescue the phenotype of N-WASP-CK2 knockdown cells, both N-WASP and
CK2 activities and abilities to assemble in a complex are required. In summary, this study shows that the
N-WASP-CK2 complex integrates in a single circuit different activities contributing to CME of EGFR and
that the interplay between the two proteins optimizes this process.
Shrew-1 wurde bei der Suche invasivitätsassoziierter Gene mittels eines DDRT-PCR-Ansatzes aus invasiven Zellen isoliert. Wie computergestützte Analysen der Sequenz ergaben, wies das bis dahin unbekannte Protein keinerlei Ähnlichkeiten mit bereits bekannten Proteinen auf und homologe Proteine wurden bisher nur in Vertebraten gefunden. Expressionsanalysen mit einem GFP-markierten shrew-1 zeigten, dass es an der basolateralen Plasmamembran lokalisiert, wo es mit dem E-Cadherin vermittelten Adhäsions-Komplex kolokalisiert. Eine Integration in diesen Komplex geschieht höchstwahrscheinlich durch direkte Interaktion mit β-Catenin. Ein weiteres Molekül das als potenzieller Interaktionspartner von shrew-1 identifiziert wurde und das in der Literatur oft als Tumorsuppressor diskutiert wird, ist Caveolin-1. Ferner konnten Überexpressionexperimente bereits zeigen, dass shrew-1 die Invasivität von HT1080-Zellen erhöhen kann. Das Ziel dieser Arbeit war es, zum einen mit Hilfe des Hefe-Split-Ubiquitin-Systems eine Interaktion von shrew-1 und Caveolin-1 zu bestätigen und zum anderen neue Interaktionspartner zu identifizieren, die helfen könnten, die Rolle von shrew-1 in invasiven Vorgängen zu erklären. Um eine mögliche Verbindung von shrew-1 und einem neuen Interaktionspartner in Bezug auf die Zellinvasivität zu untersuchen, sollten sowohl shrew-1 als auch der potenzielle Interaktionspartner mittels RNAi ausgeschaltet werden. Mit Hilfe des Split-Ubiquitin-Systems war es möglich, die Interaktion zwischen shrew-1 und caveolin-1 zu bestätigen und zu zeigen, dass diese durch die zytoplasmatische Domäne von shrew-1 vermittelt wird. Weiterhin konnte CD147 als neuer Interaktionpartner identifiziert werden. Eine Interaktion beider Proteine konnte ferner mit Hilfe des Bimolekularen-Fluoreszens-Komplementations-Systems (BIFC), des Fluoreszens-Resonanz-Energie-Transfers (FRET) und Coimmunoprezipitationen bestätigt werden. Die Interaktion von shrew-1 und CD147 scheint allerdings abhängig vom zellulären Kontext zu sein, wie die FRET-Analysen vermuten lassen. So konnte nämlich mit diesen Analysen eine starke Interaktion in MCF7-Zellen gezeigt werden, wohingegen die Interaktion in MDCK-Zellen schwächer war. Einer der auffälligsten Unterschiede dieser beiden Zelllinien im Bezug auf diese Interaktion könnte sein, dass MCF7-Zellen im Gegensatz zu MDCK-Zellen kein Caveolin-1 exprimieren. Caveolin-1 konnte seinerseits als Interaktionspartner von shrew-1 mit Hilfe des Hefe-Split-Ubiquitin-Systems bestätigt werden und andererseits wurde von einer anderen Arbeitsgruppe eine Interaktion von CD147 mit Caveolin-1 publiziert. Um dies näher zu untersuchen, wurde Caveolin-1 in MCF7-Zellen exprimiert und die FRET-Analysen in diesen wiederholt. Wie vermutet kam es zu einer Reduktion der Interaktion in Caveolin-1 exprimierenden MCF7-Zellen. CD147 ist neben vielen anderen Funktionen auch maßgeblich an der Regulation von Matrix-Metalloproteinasen beteiligt und kann somit die Invasivität von Zellen beeinflussen. Um einen Einfluß von shrew-1 und CD147 auf die Invasivität zu untersuchen, wurden beide Proteine mittels RNAi in HeLa-Zellen ausgeschaltet. Nachdem ein negativer Einfluss dieses Ansatzes auf das Proliferationsverhalten der Zellen ausgeschlossen werden konnte, wurde ein möglicher Effekt auf die Invasivität der Zellen untersucht. Durch die Analyse in Matrigel-Invasionsassays konnte gezeigt werden, dass das unabhängige Ausschalten beider Proteine die Invasivität der Zellen auf 35-55% im Vergleich zu Kontrollzellen reduziert. Die Ergebnisse dieser Arbeit untermauern die Annahme, dass shrew-1 eine Rolle bei invasiven Vorgängen spielt und weisen darauf hin, dass dies möglicherweise durch eine Interaktion mit CD147 geschieht. Die Interaktion mit CD147 und damit eine mögliche Funktion von shrew-1 bei invasiven Vorgängen scheinen dabei abhängig vom zellulären Kontext zu sein.
Batten disease refers to neuronal ceroid lipofuscinoses (NCLs), which are inherited lysosomal storage diseases with diverse ages of onset and cause progressive neurodegeneration. The most common NCL is Juvenile NCL (JNCL), which begins in early childhood and is characterized by lysosomal accumulation of subunit c of the mitochondrial ATP synthase (subunit c). JNCL is caused by mutations in the gene CLN3. This gene encodes the CLN3 protein, a transmembrane protein of unknown structure. Localization of CLN3 is ambiguous, and its exact cellular function is not known. Thereby, it is unclear what mechanisms lead to neurodegeneration in JNCL. Models of JNCL present disturbed membrane bound organelles and cytoskeleton as well as impaired autophagy and lysosomal function. The JNCL gene defect that most patients harbor is deletion of the exons 7 and 8 of CLN3. In the Cln3Δex7/8/Δex7/8 mouse model of JNCL, this deletion has been introduced to the mouse Cln3 gene.
The actin cytoskeleton consists of filaments formed through polymerization of actin and provides a framework which defines cellular morphology and also facilitates cell motility, cytokinesis, and cell surface remodeling. Rho GTPases are signaling proteins which regulate the assembly and dynamics of the actin cytoskeleton and play an important role in neuronal morphology. Rho GTPases need to be membrane-anchored in order to become active and initiate a signaling cascade. Their membrane anchorage is achieved through their geranylgeranyl tails, which they acquire through prenylation. Protein prenylation refers to the attachment of a geranylgeranyl or farnesyl group to the C-terminus of a protein. The enzyme geranylgeranyl transferase (GGTase) catalyzes geranylgeranylation, whereas geranylgeranyl pyrophosphate (GGPP) is the donor of the geranylgeranyl group. Cells produce GGPP as well as cholesterol and other lipids through the mevalonate pathway (MVA pathway).
The aim of this study was to analyze how the JNCL gene defect affects cellular morphology, especially the actin cytoskeleton and Rho GTPases, and the MVA pathway which is connected with Rho GTPase activation. These important cellular components play crucial roles in neurons and are implicated in other neurodegenerative diseases, but have received little attention in JNCL. The immortalized CbCln3Δex7/8/Δex7/8 cerebellar precursor cell line from Cln3Δex7/8/Δex7/8 mice was used for the experiments and provides a genetically accurate, neuronal cell model of JNCL. CbCln3Δex7/8/Δex7/8 cells present subunit c accumulation only when aged at confluency, but sub-confluent cells display other phenotypes. The experiments of this study were performed both with confluency-aged and sub-confluent cells. Filamentous actin was visualized, and protein levels as well as membrane localization of several small Rho GTPases was analyzed biochemically. Also the protein levels of GGTase and the key enzymes of the mevalonate pathway were determined.
Staining pattern of filamentous actin was disturbed in confluency-aged CbCln3Δex7/8/Δex7/8 cells. Additionally it was found out that these cells did not grow to wild-type size and exhibited an elongated peroxisomal morphology. Rho GTPases had reduced total levels and showed a tendency of decreased membrane localization. Levels of GGTase and the MVA pathway enzymes were altered. Results of sub-confluent CbCln3Δex7/8/Δex7/8 cells were similar with the exception of HMG-CoA reductase, which is the rate-limiting enzyme of the MVA pathway: while its level in confluency-aged CbCln3Δex7/8/Δex7/8 cells was increased, at sub-confluency it showed a reduced level. Also, in contrast with the confluency-aged cells, Rho GTPases presented a tendency of increased membrane localization.
The results of this study reveal that the accurate JNCL gene defect alters cellular morphology and the activity of the MVA pathway in neuronal cells. Small cell size and disrupted architecture of the actin cytoskeleton are confirmed as neuronal JNCL phenotypes, and the peroxisome is introduced as a novel cellular component affected in JNCL. Through defects in endocytosis, autophagy, lysosomal and mitochondrial function, and cytoskeleton, the JNCL gene defect may prevent cells from growing to wild-type size. The JNCL gene defect may attenuate the MVA pathway via mitochondrial dysfunction and/or upregulation of degradative processes. Attenuation of the MVA pathway may contribute to impaired membrane rafts, which are an established phenotype of JNCL cells. As indicated by reduced GGTase level and supported by downregulation of lipid production through the MVA pathway, the JNCL gene defect might also decrease prenylation of proteins.
Lichtsensitive Proteine bzw. Photorezeptoren eignen sich hervorragend für das Studium des Zusammenhangs von Proteinstruktur und –funktion. Lichtrezeptorproteine werden leicht durch Licht angeregt, wodurch eine gute Zeitauflösung für deren Untersuchung erreicht werden kann. Weiterhin sind sie als Signalproteine während der Etablierung des aktiven Zustandes und dessen Zerfalls großen konformationellen und strukturellen Änderungen unterworfen. Ausgehend von diesen Eigenschaften wurde bereits eine große Zahl von Lichtrezeptorproteinen genauer untersucht. Diese vorliegende Arbeit beschäftigt sich mit lichtinduzierten konformationellen Änderungen in Membranproteinen. Dafür wurden drei verschiedene Systeme herangezogen: das kleine α-helikale Peptid Gramicidin A, der G-Protein gekoppelte Rezeptor Rhodopsin and die BLUF (blue light using FAD) Domäne des hypthetischen Membranproteins Blrp (blue-light regulated phosphodiesterase) aus E. coli. Gramicidin A (gA) ist ein aus dem Bodenbakterium B. brevis isoliertes Antibiotikum, das Transportkanäle für einwertige Kationen wie Lithium, Natrium und Kalium ausbildet. Gelöst in Detergenzmizellen, wurde für gA unerwartet eine Wechselwirkung mit Blaulicht fest gestellt (Abbildung 1). Diese Beobachtung wurde mit statischen und zeitaufgelösten NMRspektroskopischen Methoden genauer untersucht und ist in Kapitel 2 näher beschrieben. Basierend auf den gewonnenen Erkenntnissen wird postuliert, dass einer der Tryptophanreste (Trp9) eine lichtinduzierte konformationelle Änderung erfährt. Ausgehend von der Konformation in Lösung befindet sich die Seitenkette von Trp9 in einem Gleichgewicht (70:30) mit einer zweiten Konformation. Bei der zweiten Konformation handelt es sich möglicherweise um die Orientierung, die der Tryptophanrest unter Festkörper-NMR Bedingungen einnimmt. Die Lebensdauer der neuen Konformation beträgt in etwa eine Sekunde. Der G-Protein gekoppelte Rezeptor Rhodopsin ist verantwortlich für die Verarbeitung von Lichtsignalen in den Stäbchenzellen der Retina. Die Absorption eines einzelnen Photons führt zur Isomerisierung des kovalent gebundenen Chromophors 11-cis-Retinal, wodurch konformationelle Änderungen im Protein veranlasst werden. Der aktivierte Metarhodopsin II (MetaII) Zustand induziert eine Enzymkaskade und schließlich einen Nervenimpuls, das Säugern das Kontrastsehen ermöglicht. Eine große Bandbreite an hochauflösenden NMRspektroskopischen Methoden, (einschließlich zeitaufgelöster und Festkörper-NMR Methoden) wurde im Laufe dieser Arbeit angewandt, um Konformation und Dynamik von bovinem Rhodopsin näher zu untersuchen. In Kapitel 3.1 sind zu Beginn mehrere Optimierungsschritte im Hinblick auf ein kostengünstiges, isotopenmarkiertes Säugerzellenmedium beschrieben. In diesem Zusammenhang wurden mehrere Rhodopsin NMR-Proben hergestellt, wobei der Gehalt an isotopenmarkierten Aminosäuren ca. 50% betrug. Anhand dieser Proben konnte bewiesen werden dass sich mit Lösungs-NMR-Spektroskopie auch sehr große, in Detergenzmizellen stabilisierte Membranproteine (~150 kD Gesamtmasse) detailliert studieren lassen. Die Untersuchungen konzentrierten sich auf den C-Terminus, für den nach sequentieller Zuordnung (Abbildung 2a) und heteronuklearern Relaxationsmessungen ein Mobilitätsverhalten bestimmt wurde, das dem mittelgroßer Proteine ähnelt. Des Weiteren konnten keinerlei definierte Strukturelemente innerhalb des C-Terminus identifiziert werden, u.a. durch einen Vergleich mit eines 19mer Peptids, dessen Primärsequenz des Rhodopsin C-Terminus entspricht (Abbildung 2a und 2b). In Kapitel 3.2 wird die nichtinvasive Zuordnung der Rückgratresonanzen aller fünf Trytophane mit Hilfe einer Kombination aus Lösungs- und Festkörper-NMR beschrieben. Dazu wurden verschiedene Rhodopsinproben hergestellt, die alle möglichen 13C’i-1-Carbonyl/15Ni-Tryptophan isotopenmarkierten Amidpaare enthielten. Eine Teilzuordnung der Tryptophanindolsignale konnte in Lösung durch Protonen-/Deuteriumaustausch und heteronukleare Relaxationsmessungen erreicht werden. Die Ergebnisse legen nahe, dass die Kombination aus Lösungs- und Festkörper-NMR-Spektroskopie sehr gut geeignet ist um komplementäre Informationen zu strukturellen und dynamischen Eigenschaften von Rhodopsin zu liefern. Fehlende Zuordnungen in den Lösungspektren konnten durch den Verglich mit Festkörperspektren ergänzt werden und umgekehrt (Abbildung 3). In Kapitel 3.3 ist die erfolgreiche Adaption der zeitaufgelösten NMR-Spektroskopie für die Untersuchung des Rhodopsin MetaII Zerfalls in vitro beschrieben. Die zeitaufgelösten protonendetektieren NMR-Experimente wurden mit unmarkiertem, in Detergenzmizellen stabilisiertem Protein bei verschiedenen Temperaturen aufgenommen, wobei sich die anschließende Auswertung auf die stark tieffeldverschobene Indolregion konzentrierte (Abbildung 4). Für die berücksichtigten Signale traten nach Induktion des aktivierten Zustandes deutliche chemische Verschiebungsänderungen auf, außerdem zeigten sie unterschiedlich schnellen MetaII Zerfall. Zusätzlich zu der erwarteten Zeitkonstante des MetaII Zerfalls (~6 min bei 298 K) konnte erstmalig eine zweite, ca. zehnmal langsamere Zeitkonstante bestimmt werden. Diese zweite Zeitkonstante ist möglicherweise ein Ausdruck für die langsame Entfaltung von Sekundärstrukturelementen nach dem Zerfall des Proteins in Opsin und Retinal. Die BLUF-Domänen verwenden Flavinadeninnukleotid (FAD) als Chromophor und gehören zu der Familie der Blaulichtrezeptoren. In Kapitel 4 wird die Untersuchung des lichtadaptierten Zustandes der E. coli BLUF Domäne auf Protein- und Ligandenebene mit zeitaufgelösten proton- und phosphordetektierten NMR-Experimenten beschrieben. In Abbildung 5 sind die statischen Licht- und Dunkelspektren (jeweils licht- und dunkeladaptiert) dargestellt. Im Folgenden konnte durch Beobachtung der Dunkeladaption bei verschiedenen Temperaturen die Aktivierungsenergie des Lichtzustandes bestimmt werden. Des Weiteren wurden zum ersten Mal phosphordetektierte NMR-Experimente erfolgreich angewandt, um einen biologisch relevanten Vorgang zeitabhängig näher zu bestimmen.
Project I: The progression of rod and cone degeneration in retinally degenerate (rd) mice ultimately results in a complete loss of photoreceptors and blindness. The inner retinal neurons survive and several recent studies using genetically targeted, light activated channels have made these neurons intrinsically light sensitive. We crossbred a transgenic mouse line expressing channelrhodopsin2 (ChR2) under the control of the Thy1 promoter with the Pde6b(rd1) mouse, a model for retinal degeneration (rd1/rd1). Approximately 30-40% of the ganglion cells of the offspring expressed ChR2. Extracellular recordings from ChR2-expressing ganglion cells in degenerated retinas revealed their intrinsic light sensitivity which was approximately 7 log U less sensitive than the scotopic threshold and approximately 2 log U less sensitive than photopic responses of normal mice. All ChR2-expressing ganglion cells were excited at light ON. The visual performance of rd1/rd1 mice and ChR2 rd1/rd1 mice was compared. Behavioral tests showed that both mouse strains had a pupil light reflex and they were able to discriminate light fields from dark fields in the visual water task. Cortical activity maps were recorded with optical imaging. The ChR2rd1/rd1 mice did not show a better visual performance than rd1/rd1 mice. In both strains the residual vision was correlated with the density of cones surviving in the peripheral retina. The expression of ChR2 under the control of the Thy1 promoter in retinal ganglion cells does not rescue vision. Project II: Lentiviral vectors are becoming the vector of choice for transgene delivery into cells due to their ability to infect non- dividing cells and stably integrate the gene into the genome of the host. Two different viral vector systems, namely HIV-1 and SIV and three different viral vectors PLECYT, PHRCMVChR2 of HIV-1 family and PBjChR2 of SIV were used in this study. The efficiency of the vectors was analyzed by applying them onto the retinal explants in culture and checking the transgene expression. The transgene in the PLECYT lentiviral vector was driven by the EF1A promoter. Upon administration of 5.2 X 106 infectious units of PLECYT viral vector suspension onto the retinal explant resulted in the transduction of retinal ganglion cells. Very few other retinal neurons were found transduced. In the case of PHRCMVChR2, approximately 5 X 105 TU/ml of the vector was used and resulted in the transduction of different neuronal subtypes. Many amacrine cells, ganglion cells and Müller cells were found expressing the transgene. For PBjChR2, 5.6 X104 TU/ml was used which resulted in Müller cell- specific transduction. Very few or no other retinal neurons were found transduced. This study demonstrates the transduction efficiency of different viral vectors on the retinal neurons in vitro. An interesting observation on these viral vectors is their altered tropism. The glycoprotein of the virus is critical for determining their tropism and in this study, all the viral vectors generated were pseudotyped with VSVG, which confers a broad non-specific spectrum of infection. However, analyzing the transgene expression, the viral vectors differ from one another and show remarkable difference in their transduction pattern. To list a few factors that might possibly responsible for the drastic transduction difference exerted by the viral vectors include; 1. Promoters used to drive the transgene expression. 2. HIV or SIV component of the vector in combination with the promoter 3. Titre of the vector used and 4. Other factors like pH and serum used in the study. Therefore optimizing the viral vectors and generating high titers would increase the efficiency and cell-type specific expression of the transgene.
5-lipoxygenase (5-LO) catalyzes the first two steps in leukotriene (LT) biosynthesis. In a two step reaction the enzyme oxygenates arachidonic acid (AA) to form the highly unstable epoxide leukotriene A4 (LTA4) in dehydrating a hydroperoxide intermediate (20). LTA4 can then be further metabolized by two terminal synthases yielding either the potent chemoattractant leukotriene B4 (LTB4) or the cysteinyl leukotrienes (CysLTs). 5-LO enzyme expression is primarily found in mature leukocytes (22) where it can either reside in the cytoplasm or in the nucleus associated with euchromatin (29). Its enzymatic activity is embedded in a complicated network in intact cells regulating LT synthesis by various factors dependent on the cell type and nature of stimulus. Factors such as the amount of free AA released by phospholipase A2 enzymes, levels of enzymes involved, catalytic activity per enzyme molecule and availability of different small molecules influence 5-LO activity (36).
The 5-LO derived LTs are lipid mediators which were shown to primarily mediate inflammatory and allergic reactions and their role in the pathogenesis of asthma is well defined. CysLTs are among the most potent bronchoconstrictors yet studied in man and play an important role in airway remodeling. LTB4 has no bronchoconstrictory effects in healthy and asthmatic humans but displays potent chemoattractant properties on neutrophils and increases leukocyte adhesion to the vessel wall endothelium (22). Therefore, LTB4 enhances the capacity of macrophages and neutrophils to ingest and kill microbes. In concert with LTB4, histamine and prostaglandin E2 (PGE2) CysLTs are thought to maintain the tone of the human airways (82).
Besides their well studied role in asthma, 5-LO derived LTs have also been implicated to play a role in cardiovascular diseases and cancer. In contrast to healthy tissues, LT pathway enzymes and receptors were found to be abundantly expressed in cancer tissues, atherosclerotic lesions in the aorta, heart and carotid artery (86). Pharmacological inhibition of 5-LO potently suppressed tumour cell growth by inducing cell cycle arrest and triggering cell death via the intrinsic apoptotic pathway (92, 93). In several studies LTs were found to exhibit cardiovascular actions by promotion of plasma leakage in postcapillary venules, coronary artery vasoconstriction and impaired ventricular contraction leading to reduced coronary blood flow and cardiac output (24). Unfortunately, the precise molecular mechanisms through which LTs influence carcinogenesis and cardiovascular diseases are still incompletely understood.
In contrast, an increasing number of studies questions the correlation between 5-LO and cancer (95-97) since extreme LT concentrations were applied to induce proliferative effects in the majority of the publications. A few studies exist which show susceptibility towards 5-LO products in physiological concentrations or achieve anti-proliferation by applying low concentrations of 5-LO inhibitors (98) ...
Juvenile Neuronal Ceroid Lipofuscinosis (JNCL) is a rare inherited childhood neurodegenerative disease that is caused by a mutation in the gene CLN3. The function of the protein produced by the gene has remained elusive, and therefore the disease mechanism of JNCL is as of yet unknown. The disease is fatal, and no cure is currently available. We believe that simvastatin shows promise as a possible treatment. Simvastatin is well tolerated in children, and as currently no other viable, less invasive treatment for JNCL exists, at least pilot-scale clinical trials for this new off-label use of simvastatin are warranted.
The protein CLN3 has been indicated to have several different subcellular localizations and functions, but conclusive evidence about its role in cellular metabolism is lacking. It is also unclear why the mutation causes the distinct phenotype of the JNCL disease. In order to bring lucidity to the issue, we set out to identify metabolic pathways related to the phenotype of JNCL by using Multi-Epitope Ligand Cartography (MELC) and the related field of toponomics. Toponomic methods are required to process the massive amount of data generated by the MELC runs in order to extract information from them.
Our disease model of choice was the CLN3Δex7/8 knock-in mouse. To separate cause from effect, we compared embryonal wild type and mutant mouse brains to their adult counterparts. The first analyses revealed progressively abnormal Combinatorial Molecular Patterns (CMPs, an unit of toponomic data) related to cholera toxin/ganglioside 1 (Ctx/GM1), which is a membrane microdomain marker.
Cholesterol is an essential part of microdomains, so we utilized filipin staining to see if there were actual changes in cholesterol concentration and localization between healthy and diseased animals. After the disturbance in cholesterol metabolism was verified, we investigated the metabolic pathway that synthesizes cholesterol, the mevalonate pathway. Simvastatin is a drug that specifically down-regulates the mevalonate pathway. Fish oil affects lipid homeostasis and has some effects similar to those of simvastatin, and both of these drugs have previously been studied for their effects on neurodegenerative diseases. After treatment of mice with these drugs, highperformance liquid chromatography (HPLC) measurements on the brain homogenate showed a decrease in levels of farnesyl pyrophosphate (FPP) and geranyl-geranyl pyrophosphate (GGPP), products of the mevalonate pathway, confirming the effect of these drugs on the brains of the animals. Analyses of motor function of the mice further supported the notion that simvastatin had a positive effect on the condition of the diseased animals.
CMP analyses from the simvastatin treated mice showed a rescue of the Ctx/GM1 CMPs, suggesting at least a partial restoration of membrane microdomain homeostasis. Filipin staining revealed reversion of the apparent cholesterol depletion in the adult mutant mouse hippocampus by simvastatin. Interestingly, an additional effect of the treatment was found: simvastatin also affected glutamate receptor homeostasis, especially as regarding to N-methyl-D-aspartate (NMDA) and alphaamino-3-hydroxyl-5-methyl-4-isoxazole-propionate (AMPA) receptors. This finding suggested that excitotoxicity could be a part of the disease process, and pointed towards glutamate receptors as possible therapy targets. This is in line with previous studies that have shown that attenuation of AMPA receptors and L voltage-dependent channels improve the phenotype of a JNCL mouse and cell model, respectively.
Simvastatin mediates many of its effects via downregulation of the mevalonate pathway products, such as isoprenoids and cholesterol. However, simvastatin also has multiple pleiotropic effects that include suppression of excitotoxicity and granting neuroprotection. It is apparent that simvastatin treatment has a positive effect on JNCL mice, but if its effects are mediated via cholesterol (and membrane microdomains), isoprenoids (and isoprenylated proteins) or via a fully cholesterol independent mechanism remains to be solved.
In this study we have shown that with the MELC method and toponomics it is possible to approach rare diseases with confounded disease mechanisms with a hypothesis-free approach, to identify possible drug targets, and to monitor the effects of the drugs on treated individuals. This should open up a new avenue in the research of the many diseases that so far have avoided all attempts at discerning their nature.
Die Etablierung eines HIV-1 Tiermodells ist ein großes Ziel auf dem Weg zur Entwicklung antiretroviraler Medikamente und Impfstoffe gegen HIV-1. Speziesspezifische Restriktionsfaktoren und fehlende Kofaktoren verhindern jedoch die Replikation von HIV-1 in Tieren. Restriktionsfaktoren sind Bestandteil der intrinsischen Immunität und entwickelten sich im Laufe der Evolution als Abwehrmechanismus gegen diverse Pathogene. Dazu gehören die Proteine der APOBEC3-Familie, TRIM5􀀁 und Tetherin, welche die Virusreplikation von HIV-1 an verschiedenen Punkten seines Lebenszyklus inhibieren. Koevolutionär entwickelten Retroviren Antagonisten, um die restriktive Funktion ihrer Wirtsproteine zu umgehen. Um ein replikationskompetentes, simiantropes HIV zu generieren, wurden im Rahmen dieser Arbeit die Sequenzen vifHIV-1 und vpuHIV-1 gegen vifagm.tan aus SIVagm.tan und vpugsn/den aus den Immundefizienzviren SIVgsn und SIVden substituiert, um die Restriktion gegen A3G und Tetherin in Zellen der Afrikanischen Grünen Meerkatze zu umgehen. Die TRIM5 vermittelte Restriktion wurde über eine Mutation in der Cyclophilin A Bindedomäne des Kapsids verhindert. Die Analyse der Vifagm.tan Funktion bestätigte den geänderten Tropismus des chimären HIV-1 bezüglich der APOBEC3G vermittelten Restriktion. Denn nach Austausch des vifHIV-1 Gens war das Virus nicht mehr in der Lage, die Aktivität des humanen APOBEC3G zu unterbinden und initiierte stattdessen die Degradation des Analogons aus der Afrikanischen Grünen Meerkatze. Weiterhin konnte die erfolgreiche Klonierung des vpugsn/den Gens in HIV-1 die Aktivität der SHIV-Konstrukte gegen Tetherin der Afrikanischen Grünen Meerkatze und der Rhesusaffen ändern, wohingegen humanes Tetherin nicht mehr abgebaut werden konnte. Trotz der erfolgreichen Aktivität der konstruierten SHIVs gegen die zellulären Restriktionsfaktoren der Afrikanischen Grünen Meerkatze, replizierten die generierten chimären Viren in einer AGM Zelllinie, nicht aber in periphären mononuklearen Blutzellen der Afrikanischen Grünen Meerkatze. Ein Indiz, das für weitere strukturelle Anpassungen der Viren gegenüber ihren Wirten spricht, die zur Bildung der Spezies-Barriere beitragen und Zoonosen erschweren. Im zweiten Teil der Dissertation wurden die Aktivitäten der Proteine der APOBEC3-Familie und VifHIV-1 auf ihre Regulation durch Phosphorylierung untersucht. Proteinphosphorylierungen gehören zu den wichtigsten posttranslationalen Proteinmodifikationen, um diverse Funktionen wie die Enzymaktivität, Proteininteraktionen und die zelluläre Lokalisation zu steuern. Dabei konnte die durch Yang et al. postulierte Phosphorylierung von VifHIV-1 nicht bestätigt werden. Analysen der mutmaßlichen VifHIV-1 Phosphomutanten enthüllten, dass die Funktion von VifHIV-1, die Infektiosität von HIV-1 zu gewährleisten, durch Substitution der mutmaßlichen Phosphoaminosäuren nicht beeinträchtigt wird und ebenso sämtliche Phosphomutanten die Degradation von A3G initiierten, wenn auch in unterschiedlichem Maße. Weiterhin wurde im Rahmen dieser Arbeit gezeigt, dass A3C entweder in einem phosphorylierten Protein-Komplex vorliegt oder ein phosphoryliertes Protein bindet. Zudem konnte ermittelt werden, dass APOBEC3A als einziges Protein der APOBEC3-Familie nach TPA- und cAMP-Stimulation phosphoryliert wird. In vitro Kinase Studien konnten zeigen, dass die Phosphorylierung unter anderem durch ERK2 erfolgt. Es konnte jedoch kein Zusammenhang zwischen der Phosphorylierung von A3A und dessen zellulärer Lokalisation, Aktivität gegen HIV-1 als auch gegen die Retrotranspositionselemente IAP und LINE-1 hergestellt werden. Dies lässt den Schluss zu, dass die Phosphorylierung die untersuchten Aktivitäten von APOBEC3A nicht beeinflusst oder APOBEC3A eine bisher unbekannte durch Phosphorylierung regulierte Funktion besitzt.
Im Rahmen dieser Arbeit sollte ein Anti-PGE2-Antikörperfragment exprimiert werden, mit dem über NMR-Spektroskopie genauere Strukturdaten der Antikörperbindungsdomäne zu erhalten wären. Da Fab-, Fv- und scFv-Fragmente die Antigenbindungsstelle besitzen und sich gegenüber einem vollständigem IgG zusätzlich durch ihre geringere Größe auszeichnen, wurden die korrespondierenden Antikörperfragmente für diese Arbeiten gewählt. Ausgehend von einer etablierten Hybridom-Zelllinie konnten über cDNA-Synthese und Assembly-PCR die Gene für die Antikörperfragmente bereitgestellt werden. Zum Verifizieren von DNA-Sequenz und Bindungseigenschaften stand ein Anti-PGE2-Fab-Fragment früherer Arbeiten von Frau Dr. Ilse Zündorf zur Verfügung. Für Strukturuntersuchungen müssen relativ große Proteinmengen bereit gestellt werden, daher ist es notwendig, hohe Ausbeuteverluste durch Präzipitation, Degradierung, Aufreinigungsverluste u.a. zu vermeiden. Um zunächst eine möglichst gute Proteinexpression zu erhalten, wurde der Bakterienstamm Escherichia coli BL21DE3 und das pET-Expressionssystem gewählt. In diesem System war zu erwarten, dass die überexprimierten Antikörperfragmente unter den gewählten Bedingungen in Form unlöslicher Aggregate, sogenannter inclusion bodies, in den Zellen vorliegen werden. Die der Klonierung der VH-, Vk- und scFv-Gene folgende Expression der Proteine zeigte, dass es tatsächlich zur Bildung von inclusion bodies kommt. Die Vorteile dieser Aggregatbildung, wie hohe Proteinmenge, Proteaseunempfindlichkeit aufgrund der hohen Dichte sowie die hohe Homogenität, sollten für die Gewinnung nativer Antikörperfragmente genutzt werden. Hierfür konnte ein System etabliert werden, das im wesentlichen auf der Solubilisierung der Aggregate und der Reduktion falscher Disulfidbrücken mit anschließender Rückfaltung und Rekonstituierung der Disulfidbrücken beruht. Kritische Parameter, die den Rückfaltungsprozess beeinflussen, wurden untersucht. Hierunter fallen insbesondere die Zusammensetzung des Renaturierungssystems wie Pufferbedingungen, das Verhältnis von oxidierten zu reduzierten Glutathion und der Zusatz von sogenannten Faltungsenhancern, die Additiva. Für fast alle wesentlichen Punkte konnten Systemkomponenten etabliert werden, die für das scFv-Fragment eine nahezu vollständige Überführung der inclusion bodies in die lösliche Form erlauben. Auch eine Bindung an das Antigen Prostaglandin E2 konnte unter Anwendung von Radioimmunoassays gezeigt werden. Problematisch ist die Aufkonzentrierung der in niedriger Konzentration vorliegenden renaturierten scFv-Fragmente. Je stärker aufkonzentriert wird, desto stärker präzipitieren die scFv-Moleküle in der Ultrafiltrations- bzw. Zentrifugationsfiltereinheit. Antikörper und Antikörperframente sind ohne Frage eine sehr wichtige Molekülgruppe von molekularbiologischen und biochemischen Interesse, nicht nur in dieser Arbeitsgruppe. Im Rahmen einer möglichen Weiterführung dieser Arbeit müsste geklärt werden, ob das Präzipitationsverhalten der scFv-Fragmente eine inhärente Eigenschaft des Anti-PGE2 scFv ist, oder ob hier doch eine zumindest teilweise inkorrekte Faltung der Proteinkette vorliegt. Mögliche weitere Experimente könnten in der Variation des Linkers liegen oder das etablierte Verfahren an einem weiteren scFv-Fragment zu testen. Da die genannten Probleme bei der Aufkonzentrierung der Proteinlösung auftreten, sollten statt der angewandten Techniken alternative Methoden geprüft werden. Auch wäre es interessant zu sehen, ob sich ein z.B. kommerziell verfügbares scFv-Fragment, dessen Bindungseigenschaften bekannt sind, nach Denaturierung wieder in die native Form überführen lässt. Abschließend lässt sich sagen, dass die Herstellung von nativen Protein aus inclusion bodies viele Vorteile bietet, die für die Präparation größerer Proteinmengen nicht ungenutzt bleiben sollten.
Parvulustat ist ein kleines Protein (8,2 kDA), das aus dem Bodenbakterium Streptomyces
parvulus sekretiert wird. Es ist ein saures Polypeptid mit einem isoelektrischen Punkt von 4,49
und inhibiert spezifisch α-Amylasen (Ki = 9 x 10-12).
In der vorliegenden Arbeit sollen neben der Isotopenmarkierung des Parvulustats PL-Mutanten
mittels PCR-Mutagenese und DNA Rekombinationstechnick hergestellt werden.
Zur Charakterisierung und Strukturbestimmung der Mutanten werden größere Mengen des
jeweiligen Proteins benötigt, wofür wiederum ein effektives Expressionssystem bereitstehen
muß. Ein Expressionssystem besteht aus einem Wirtsorganismus der seinen Protein-Biosynthese-
Apparat zur Verfügung stellt und einem Vektor (Plasmid) der das Zielgen trägt. Im Falle des
Parvulustats und seiner Mutanten bietet sich der gut charakterisierte Stamm Streptomyces
lividans TK 24 (Hopwood et al, 1985) als Wirtsorganismus an. Der Inhibitor wird in ihm als
Vorläuferprotein mit Signalpeptid als Transportsignal (Leader) gebildet. Da dieses Bakterium
keine äußere Membran besitzt, wird der Inhibitor direkt ins Kulturmedium ausgeschleust. Dabei
wird das Signalpeptid durch die Signalpeptidase abgespalten.
Das Strukturgen wird zuerst im Klonierungsplasmid pSH09 in E. coli vervielfältigt, bevor es mit
den Enzymen EcoRI und SpeI geschnitten wird, um in den Expressionsvektor pSH017 integriert
zu werden. Dieser Shuttlevektor trägt genauso die gewünschten Restriktionsschnittstellen, SpeI
und EcoRI, die die paßgenaue Ligation des rekombinanten Strukturgens mit den funktionellen
Elementen erlauben.
Nach erfolgtem Einbau des Zielgens in das neue Vektorkonstrukt kann dieser Expressionsvektor
leicht in den Wirtsorganismus eingebracht werden und sich dort vermehren. Nach Klonierung der
mutierten Parvulustatsgene in den Expressionsvektor werden die damit transformierten Zellen auf
R2YE-Platten aufgebracht und bei einer Temperatur zwischen 20 und 28°C je nach PL-Mutanten
inkubiert. Zur Selektion wird Thiostrepton verwendet. Nach Transformation und Vermehrung
geeigneter Wirtszellen wird das klonierte Gen transkribiert. Durch anschließende Translation der
gebildeten mRNA in der Wirtszelle entsteht das gewünschte Protein in großen Mengen. Zur
Überprüfung der Aktivität der einzelnen Mutanten steht der qualitative α-Amylase-Plattentest zur
Verfügung, wobei die aktiven Mutanten einen blauen Hemmhof bilden (siehe Abbildung 24). Der
blaue Jod-Stärke-Komplex weist, bei gleichzeitigem Vorhandensein von α-Amylase, auf nicht
abgebaute Stärke und damit indirekt auf die Produktion des α-Amylase-Inhibitors hin.
Da das Parvulustat von den Bakterien ins Medium abgegeben wird, besteht die Abtrennung der
Zellen durch Zentrifugation des Kulturmediums. Es folgen Ammoniumsulfatfällung des
Überstandes, Auftragung auf PAGE und präparative HPLC, nach der reines, einheitliches Protein
isoliert werden konnte.
Kollagen des marinen Schwammes Chondrosia reniformis Nardo: Optimierte großtechnische Herstellung, Analytik und neue pharmazeutisch-technologische Anwendungsmöglichkeiten Großtechnische Isolierung und Untersuchung von Chondrosia-Kollagen Schwämme spielen eine wichtige Rolle im Ökosystem des Meeres und ihr Vorkommen ist begrenzt. Zur schonenden und nachhaltigen Nutzung als Rohstoffquelle müssen im Hinblick auf eine industrielle Nutzung alternative Wege zur Gewinnung von marinem Schwammkollagen entwickelt werden. Ein Beispiel hierfür sind die sogenannten „Marikulturen“ direkt im Meer, aus denen die im Rahmen dieser Arbeit verwendeten Schwämme stammten. Im Gegensatz zu den früher eingesetzten Methoden zur Isolierung des Chondrosia-Kollagens, die Laborcharakter trugen, konnte in der vorliegenden Dissertationsarbeit ein stark vereinfachtes, für die großtechnische Produktion geeignetes Verfahren mit einer hohen Ausbeute (ca. 35%) beschrieben werden. Das Schwammausgangsmaterial wurde über ca. 4 Tage mit Hilfe von Natriumsulfit reduktiv behandelt und die Kollagenfasern (nach einem Filtrationsschritt) in saurem Medium gewonnen. Atomkraftmikroskopische Aufnahmen des isolierten Materials zeigten die kollagentypische Fibrillenquerstreifung, die Unlöslichkeit der Faserbündel in saurem Milieu sowie deren Abbau in die einzelnen Fibrillen in neutraler Pufferlösung. Die von tierischen Kollagenen abgeleitete Bestimmung des Proteingehaltes ergab einen im Vergleich zum kalkulierten Kollagenanteil höheren Gesamtproteingehalt. Dies ist ein Hinweis auf mögliche nichtkollagene Proteinanteile, die an Kollagen assoziiert vorliegen können. Untersuchungen zur Aminosäurenzusammensetzung des isolierten Materials ergaben im Vergleich zu den von Swatschek et al. (2002a) publizierten Ergebnissen einen deutlich höheren Glycin- und einen mehr als doppelt so hohen Hydroxyprolingehalt. Dies kann dahingehend gedeutet werden, dass die neue Isolierungsmethode die bekannten Verunreinigungen des Kollagens mit Glykoproteinen reduziert und zu einem deutlich reineren Kollagenmaterial führt. Mit Hilfe einer neu entwickelten Methode konnten erstmalig Partikel aus Chondrosia-Kollagen hergestellt werden, deren Größe im Nanometerbereich lag. Hierbei handelte es sich um eine kontrollierte alkalische Hydrolyse des isolierten, gefriergetrockneten Kollagens in 1,5 M NaOH. Es konnte eine Partikelausbeute in Höhe von ca. 10% erzielt werden. Atomkraftmikroskopische Aufnahmen zeigten gleichförmige, sphärische Partikel. Die Partikelgrößenuntersuchungen mittels Photonenkorrelationsspektroskopie (nach Resuspension der gefriergetrockneten Schwammkollagennanopartikel (SKNP)) ergaben eine Größe von 168 ± 9,1 nm. Somit sind diese Partikel im Gegensatz zu den bisher aus Chondrosia-Kollagen gewonnenen Mikropartikeln auch für intravenöse Anwendungen geeignet und haben eine größere Trägerkapazität. Die hergestellten Nanopartikel wurden adsorptiv mit dem Wirkstoff 17?-Estradiol-Hemihydrat beladen. In Abhängigkeit von der eingesetzten Stoffkonzentration (1,25 bis 5,0 mg/ml) konnten Beladungsraten von bis zu 13,1% erzielt werden. Die Stabilitätsprüfung von wässrigen Partikelsuspensionen ergab nach 180 Tagen Einlagerung eine auf 7 bis 8% reduzierte Beladungsrate. Nach Inkorporation der beladenen Partikel in eine Hydrogelgrundlage wurde die transdermale Bioverfügbarkeit des Arzneistoffs im Rahmen der Hormonersatztherapie bei postmenopausalen Frauen untersucht. SKNP-Gele mit einer Estradiolkonzentration in Höhe von 0,06% wurden mit gleichkonzentrierten partikelfreien Gelen verglichen. In den Speichelproben der Patientinnen konnten nach täglicher einmaliger Applikation des Nanopartikelgels nach 28 Tagen signifikant höhere Estradiolkonzentrationen gemessen werden. Die ermittelten Flächen unter den Konzentrations-Zeit-Kurven (AUC) über 24 h nach der einmaligen topischen Applikation von 0,75 mg Estradiol waren um das 2,3 bis 3,4-fache höher als die entsprechenden AUC-Werte des Vergleichsgels. 24 h nach Auftragen des Gels lagen die Estradiolspiegel des Vergleichsgels in etwa wieder auf Höhe der Basalwerte, wohingegen die Estradiolwerte nach Applikation des SKNP-Gels zum gleichen Messzeitpunkt mindestens doppelt so hoch waren. Somit konnten bei Anwendung der SKNP-Zubereitung eine deutlich erhöhte transdermale Estradiolabsorption und eine verlängerte Arzneistoffwirkung beobachtet werden. Das nanopartikuläre Trägersystem aus Chondrosia-Kollagen stellt damit eine vielversprechende Möglichkeit zur Erhöhung der transdermalen Bioverfügbarkeit von lipophilen, schlecht resorbierbaren Wirkstoffen dar. Im Rahmen der vorliegenden Arbeit wurde eine Formulierung für einen magensaftresistenten Überzug mit Chondrosia-Kollagen als Filmbildner entwickelt. Die Sprühsuspension bestand aus 15% gefriergetrocknetem Kollagen, dem Weichmacher Triethylcitrat (1,5%), dem Trennmittel Talkum (7,5%) und Wasser. Der Coating-Prozess wurde in einem Dragierkessel mit Placebotabletten durchgeführt. In regelmäßigen Abständen wurden Tabletten-Stichproben entnommen, um die erforderliche Kollagenschichtdicke für eine Magensaftresistenz zu bestimmen. Tabletten mit einer Kollagenauftragsmenge von 12,9 mg/cm2 entsprachen der Prüfung auf Magensaftresistenz gemäß Europäischem Arzneibuch. Die Widerstandsdauer in 0,1 M Salzsäure betrug mehr als 2 h und der Zerfall in Phosphatpufferlösung pH 6,8 höchstens 10 min. Rasterelektronenmikrosko-pische Aufnahmen bei unterschiedlichen Vergrößerungen zeigten eine gleichmäßige und glatte beschichtete Oberfläche. Das Auftragsverfahren im Dragierkessel erwies sich als reproduzierbar. Die mechanischen Eigenschaften der überzogenen Tabletten waren zufriedenstellend und die Magensaftresistenz wurde durch eine 6-monatige Lagerung nicht beeinträchtigt. Als Naturprodukt eignet sich Schwammkollagen als Überzugsmaterial für Nichtarzneimittel. Der schnelle Zerfall des Kollagenüberzugs in neutraler Pufferlösung stellt einen Vorteil gegenüber den häufig verwendeten Schellacküberzügen dar, die sich im Darmtrakt zu langsam auflösen und daher nicht selten mit synthetischen Polymeren kombiniert werden.
Das humane MLL-Gen (Mixed Lineage Leukemia) ist in chromosomale Translokationen mit mehr als 50 verschiedenen Partnergenen involviert. Alle diese Rearrangements sind mit akuter lymphatischer oder akuter myeloischer Leukämie assoziiert. Die reziproke Translokation t(4;11) ist ursächlich für die Entstehung einer akuten lymphatischen Leukämie, die gehäuft bei Kleinkindern auftritt. Die Prognose dieser aggressiven Leukämie ist sehr schlecht, da die leukämischen Blasten nahezu Therapie-resistent sind. Für einige MLL-Rearrangements konnte in Transduktions/Transplantations-Experimenten gezeigt werden, dass das MLL-Fusionsgen akute myeloische Leukämien in Mäusen induzieren kann. Aus diesem Grund steht immer noch das jeweilige MLL-Fusionsprotein im Mittelpunkt der Suche nach einem zugrunde liegenden Pathomechanismus. Die Versuche, ein Tiermodell mit dem MLL/AF4- Fusionsprodukt zu etablieren, blieben lange Zeit erfolglos. Erst Anfang dieses Jahres gelang es, durch eine knock-in Strategie, transgene Mäuse zu generieren, die das MLL/AF4 Translokationsprodukt tragen. Diese Mäuse entwickeln nach einer sehr langen Latenzzeit von bis zu 540 Tagen diffuse B-Zell-Lymphome. An der t(4;11) sind die Gene MLL auf Chromosom 11q23 und AF4 auf Chromosom 4q21 beteiligt. Die reziproke Translokation führt zur Entstehung von zwei Fusionsgenen (MLL/AF4 und AF4/MLL) mit intaktem Leserahmen. Wir nehmen an, dass beide Fusionsgene notwendig sind, um die Entartung der Zellen zu bewirken. Zum Einen ist der beschriebene Phänotyp der transgenen MLL/AF4 positiven Mäuse mit dem aggressiven Verlauf der t(4;11) Leukämie nicht vergleichbar, und zum Anderen findet man bei der Mehrzahl der Patienten mit einer t(4;11) beide Fusionstranskripte in den leukämischen Blasten. Um die Eigenschaften der Fusionsproteine AF4/MLL und MLL/AF4 zu untersuchen, wurden stabil transfizierte murine embryonale Fibroblasten-Linien etabliert, die entweder eines oder beide Fusionsgene regulierbar exprimierten. Mit Hilfe dieses in vitro Modells konnte man erstmals die Auswirkungen der gleichzeitigen Expression beider Fusionsgene auf das Verhalten der Zellen analysieren. Die Untersuchungen der Proliferation und der Apoptoseraten der verschiedenen transfizierten Zelllinien ergab, dass das AF4/MLL Fusionsprotein in den Zellen zu Wachstumstransformation führt, aber gleichzeitig die Zellen für den Zelltod durch bislang unbekannte Apoptosemechanismen sensitiviert. Das MLL/AF4 Translokationsprodukt war nicht in der Lage, eine Wachstumstransformation auszulösen. Vielmehr wurde deutlich, dass die Expression von MLL/AF4 die Proliferation der Zellen hemmt. Die Untersuchung der spezifischen Apoptoserate in den MLL/AF4 positiven Zellen ergab, dass die Zellen sowohl unter normalen Bedingungen als auch unter Stress-Konditionen Apoptose-resistent waren. Damit besitzen beide Fusionsproteine onkogene Eigenschaften. Da nur die mit AF4/MLL und MLL/AF4 transfizierten Zellen eine stark erhöhte Proliferation aufwiesen, im Wachstum transformiert waren und eine erhöhte Apoptoseresistenz zeigten, wurde deutlich, dass die Eigenschaften beider Fusionsproteine notwendig sind, um den vollständig transformierten Phänotyp auszulösen. Um die Veränderungen in den Zellen auf molekularer Ebene zu untersuchen, wurde mittels quantitativer PCR die Expression verschiedener Zellzyklus- und Apoptose-regulierender Gene untersucht. Dabei wurde deutlich, dass die Expression Zellzyklus- und Apoptose-regulierender Gene in allen drei Zellen verändert war. Genexpressionsprofil-Studien der Zellen ergaben schließlich, dass der Transkriptionsfaktor Nanog in den doppelt-transfizierten Zellen verstärkt exprimiert wurde. Nanog ist in die Aufrechterhaltung der Pluripotenz und des Selbsterneuerungspotenzials von embryonalen Stammzellen involviert. In dieser Arbeit konnte erstmals der Phänotyp einer Zelllinie beschrieben werden, die beide Fusionskonstrukte, AF4/MLL und MLL/AF4, stabil exprimierte. Dabei wurde deutlich, dass nur durch die Expression beider Fusionsgene ein Phänotyp erzeugt wird, der den Leukämoblasten sehr ähnlich ist. Die Expression von Nanog in diesen Zellen erklärt den Stammzell-Charakter der leukämischen Zellen.

Particular interest in photosynthesis research is focused on diatoms and as one of the major players of marine phytoplankton, diatoms have a huge impact on global photosynthesis.
Diatoms originated from a secondary endosymbiosis that took place between a putative photosynthetic red algal ancestor and a heterotrophic eukaryote. Secondary endosymbiosis resulted in the formation of chloroplasts with four membranes. Centric diatoms (e.g. Thalassiosira pseudonana or Cyclotella meneghiniana) usually possess many small chloroplasts, while pennates (e.g. Phaeodactylum tricornutum) have several larger ones, or even only one which can occupy half of the cell volume...
5-lipoxygenase (5-LO) is an enzyme with a substantial role in inflammatory processes. In vitro kinase assays using [32P]-ATP in combination with mutagenesis have revealed that serine residues 271, 523 and 663 can be phosphorylated by MK2, PKA and ERK2 kinases, respectively. A few available reports regarding 5-LO protein sequence have covered up to 30% of the sequence after amino acid sequencing including Ser663. In LCMS/MS analyses of 5-LO tryptic digests from different cellular sources different peptides have been detected; however, none of the three phosphorylations has been detected and only Ser663 was included in the covered sequence.
As there was no comprehensive mass spectrometric analysis of 5-LO, the purpose of this study was to optimize the experimental conditions under which detection of the aforementioned phosphorylation events, as well as other possible post-translational modifications (PTMs), would be feasible. Matrix-Assisted Laser Desorption/Ionization Mass Spectrometry (MALDI-MS) was used for peptide analysis of 5-LO cleaved either by chemical reagents or by proteases. Sequence coverage of 5-LO could be enhanced to be close to completion by combination of results from digestions by trypsin, AspN and chymotrypsin. In-gel trypsin digestion followed by in-solution AspN digestion proved to be a useful sample treatment for reproducible detection of the Ser271-containing peptide.
Nevertheless, in none of the examined cleavage protocols the sequence around Ser523 was detected reproducibly or with acceptable signal intensity for subsequent peptide fragmentation. Propionic anhydride and sulfo-NHS-SS-biotin cross-linker (EZ-linkTM), were used for derivatization of lysine side chains and hindrance of lysine residue recognition by trypsin. Phosphopeptide enrichment became possible after tryptic digestion of these samples, not only due to formation of an individual Ser523-containing peptide, but also because TiO2-mediated enrichment, which is performed in acidic pH, was not impaired by positively charged free lysine side chains. Additionally, biotinylation of lysine residues was exploited for an intermediate enrichment step of the lysine containing peptides, prior to TiO2 phosphopeptide enrichment.
MALDI-MS analysis after in-vitro phosphorylation of 5-LO by the three kinases showed that Ser271 was phosphorylated in the MK2 and PKA kinase assays, while Ser523 was phosphorylated only in the PKA kinase assay. Surpisingly, no phosphopeptides were detected in the in-vitro kinase assays with ERK2, even though the unmodified counterpart of the Ser663-containing peptide was easily detected. The detection limit for each of the three phosphorylation sites was determined by the use of custom made phosphopeptides and an amount of 0.06 pmol of phosphopeptide in 1 μg 5-LO (representing 0.5% phosphorylation rate) was sufficient in all cases for successful enrichment and detection by MS.
In-vitro kinase assays with [32P]-ATP were performed for some kinases that were expected to phosphorylate 5-LO according to in-silico data. Three members of the Src tyrosine kinase family (Fgr, Hck and Yes) and the Ser/Thr specific kinase DNA-PK used 5-LO as their substrate and mainly residues at the N-terminal part of 5-LO were detected phosphorylated by MS (e.g. Y42, Y53). Additional in-vitro assays for recombinant 5-LO modification included incubation with glutathione or compound U73122, previously described as inhibitor of 5-LO.
Since in-vitro assays might have generated artifacts, a method for 5-LO purification from human cells was sought, in order to examine the modification state of the protein in the cellular context. ATP-agarose affinity purification and anti-5-LO immunoprecipitation proved inappropriate for sample purification for MALDI-MS analysis. Consequently, two human cell lines that are able to express 5-LO (Rec-1 Blymphocytes and MM6 monocytes) were transduced with a DNA cassette that contained recombinant human 5-LO sequence with an attached N-terminal FLAG-tag. Anti-FLAG immunoprecipitation was then performed effectively in cell lysates and the precipitated FLAG-5-LO was separated by SDS-PAGE before MALDI-MS analysis.
The examined cell stimuli were expected to result to phosphorylation of 5-LO at Ser523 by PKA in Rec-1 cells and to phosphorylation of Ser271 and/or Ser663 in MM6 cells by activated MK2 and ERK2, respectively. Additionally, under the conditions of MM6 cell stimulation, Fgr, Hck and Yes kinases, which phosphorylated 5-LO in vitro, were expected to be activated and the possibility of 5-LO phosphorylation on tyrosine was investigated. Although immunoblotting results indicated that all the aforementioned phosphorylation events existed in the examined samples, MALDI-MS analysis verified only phosphorylation on Ser271 in differentiated MM6 cells, interestingly regardless of cell stimulation.
Finally, the primary amine derivatization procedure by EZ-linkTM was utilized for MS analysis of lysine rich proteins. In the past, chemical propionylation of histones had been employed prior to trypsin digestion; however it was easily confused in MS with combinations of other PTMs (e.g. acetylation, methylation). Moreover, propionylation is a PTM for histone H3 and this information was lost. Consequently, the EZ-link reagent was more useful for analysis of histones, as unambiguous assignment of PTMs and detection of native propionylation on bovine H3 became possible.
Die Brustdrüse (glandula mammaris) bietet ein einzigartiges Modellsystem zum Studium der adulten Stammzellen und der molekularen Signalwege, welche die Selbsterneuerung dieserZellen sowie die Proliferation und Differenzierung der Vorläuferzellen kontrollieren. Die Brustdrüse besteht aus dem Brustepithel und dem Stroma, das zum größten Teil aus dem Fettgewebe gebaut ist. Es enthält auch andere Zelltypen z. B. Fibroblasten und Makrophagen. Die Entwicklung der Brustdrüse findet hauptsächlich nach der Geburt, während der Pubertät, Schwangerschaft und Laktation statt. Ein funktionelles Brustepithel wird während der aufeinander folgenden Zyklen von Schwangerschaft, Laktation und Abstillen auf- und wieder abgebaut. Diese Regenerations-Kapazität kann für die Organrekonstitution genutzt werden. Die Transplantation der kleinen Anzahl von Brustepithelzellen oder des Drüsenfragments in das Fettgewebe einer Empfängermaus, deren eigenes Brustepithel entfernt wurde (cleared fat pad), führt zur vollständigen Epithelregeneration. Die zyklische Entwicklung und Regenerations-Fähigkeit des Epithelgewebes lässt auf die Existenz von Stammzellen schließen, die im Verbund der Epithelzellen überdauern. Diese gewebespezifischen Stammzellen sind in der Lage sich durch asymmetrische Zellteilung zu erneuern (self-renewal) und gleichzeitig die differenzierenden Vorläuferzellen zu bilden. Die während der Pubertät und Schwangerschaft erhöhten systemischen Hormone, lokalen Wachstumsfaktoren und Zytokine kontrollieren die Stammzellen-Proliferation und die Differenzierung der Vorläuferzellen in den verschiedenen Brustepithel-Zelllinien: Myoepithel-, Luminal- und Alveolarzellen. Aufgrund der Tatsache, dass die Entstehung von Brustkrebs mit aberranten Proliferations- und Differenzierungsprogrammen in malignanten Stamm-/ Vorläuferzellen (cancer stem cells) einhergeht, ist die Identifizierung der Signalwege, die diese Prozesse regulieren, für die Stammzellen- und Krebs-Forschung sehr bedeutend. Basierend auf diesen Erkenntnissen wurden im Rahmen des vorliegenden Projektes die Methoden zur genetischen Manipulation von nicht-angereicherten Brustdrüsen-Stammzellen entwickelt. Durch effiziente lentivirale Transduktion von adhärenten Primärzellen wurden nahezu 90% der Zellen, einschließlich der Stammzellen, transduziert. Diese Optimierung erfolgte durch 1) die Anwendung von konzentrierten Lentiviren mit hoher Qualität, 2) Passagierung der Primärzellen und Entfernung von Gewebeklumpen von den VIII Primärzellkulturen, und besonders 3) durch die Reduzierung der Zelldichte während der viralen Transduktion. Für Brust-Stammzellen sind keine spezifischen Oberflächen-Marker bekannt und daher ist ihre Isolierung deutlich erschwert. Man konnte sie bis jetzt nur anhand der moderaten Expression von CD24 (hitzestabilen Antigen) und hoher Expression von CD49f oder CD29 (α6- oder β1-Integrin) ungefähr 10-fach anreichern. Allerdings haben andere Studien gezeigt, dass die Transplantation der FACS-sortierten Stammzellen zu einer Schädigung der Stammzellen und folglich zu einer Reduktion der Repopulation-Frequenz führen kann. Aus diesem Grund wurden die genetisch modifizierten Stammzellen nicht sortiert. Durch die Transplantationen der transduzierten Primärzellen wurde ihr Stammzellen-Anteil in ihrer natürlichen Nische (cleared fat pad) selektiert. Die transplantierten Stammzellen sind in der Lage duktale Auswüchse zu entwickeln. Mit dieser Strategie konnten Transplantate mit homogener Expression von Fluoreszenz-Markergenen, wie z. B. GFP, erzielt werden. FACS Analysen der Zellen, die aus Transplantaten isoliert werden, haben gezeigt, dass alle drei Brustepithelzell-Populationen, nämlich Luminal-, Basal- und Stammzellen, transduziert waren und GFP exprimierten und daher aus transduzierten Zellen hervor gingen. Die Transplantationen einer Mischung der unterschiedlich fluoreszenzmarkierten Stammzellen ergaben einzelne verzweigte Auswüchse, in denen jeweils nur ein Fluoreszenz-Markerprotein exprimiert wurde. Sie stammen sehr wahrscheinlich von einzelnen transduzierten Stammzellen ab und wachsen jeweils in einem begrenzten Bereich des Brustfettgewebes aus. Die Immun-Antwort der Empfängermäuse gegen Fluoreszenz- Markerproteine könnte das Auswachsen der Transplantate inhibieren. Brustepithelium-Rekonstitutionen waren daher in den Rag2-/-γc-/- Empfängermäusen mit geschwächtem Immunsystem besonders effiziert. Die lentivirale Manipulation von Stammzellen und deren Einsatz in Brustepithelium-Rekonstitutionen kann als alternative Methode zur gewebsspezifischen Knockout-Technik angesehen werden. Für die Etablierung dieser Methode wurde im Rahmen dieser Arbeit ein zentraler Transkriptionsfaktor in der Brustentwicklung, signal transducer and activator of transcription 5 (Stat5), untersucht. Massenspektrometrische Untersuchungen von DNA, RNA und nichtkovalenten Oligonukleotid-Komplexen

Ziel dieser Dissertation war es, grundlegende Untersuchungen zum massenspektrometrischen Nachweis von DNA und RNA durchzuführen, sowie der spezifische Nachweis von nichtkovalenten Oligonukleotid-Komplexen. Dabei lag der Schwerpunkt auf den folgenden drei Bereichen: - Allgemeiner Vergleich zwischen DNA und RNA sowie den Ionisierungsmethoden nano-ESI und MALDI - Quantitative Untersuchung des Einflusses des Salzgehaltes und der Aufreinigung der Probe auf das Spektrum - Grundlegende Vorraussetzungen zum spezifischen Nachweis nichtkovalenter Komplexe mittels Massenspektrometrie (MS) Dabei zeigte sich beim massenspektrometrischen Vergleich von DNA mit RNA, dass die RNA stets eine höhere Stabilität aufweist, wobei diese bei beiden Oligonukleotiden mit steigender Kettenlänge abnimmt. Gleichzeitig erhöht sich mit steigender Kettenlänge die Tendenz zur Kationenanlagerung im Spektrum. Um diese Anlagerungen zu unterdrücken, ist die Verwendung von ammoniumhaltigen Zusätzen ist bei der MS stets erforderlich. Bei MALDI haben die Matrices einen großen Einfluss auf das Fragmentierungsverhalten der Proben. Für die Oligonukleotide konnte daher folgende Einteilung der Matrices von „hart“ nach „weich“ getroffen werden: HCCA > DHB > THAP ungefähr gleich ATT > HPA. Bei der Untersuchung von Oligonukleotiden mittels nano-ESI konnte gezeigt werden, dass die Verwendung von Quarzkapillaren der Verwendung von Glaskapillaren vorzuziehen ist, da mit Quarz die Anlagerungen der Kationen im Spektrum deutlich unterdrückt werden. Weiterhin wurde festgestellt, dass die Erhöhung des Drucks in der ersten Druckstufe zu einer schonenderen Desolvatisierung und damit zu einer deutlich verbesserten Nachweisgrenze führt. Bei der quantitativen Untersuchung des Salzeinflusses konnte eine maximale Salzkonzentration ermittelt werden, bei der eine massenspektrometrische Untersuchung von DNA und RNA noch möglich ist. Dabei hat sich gezeigt, dass bei direktem Vergleich nano-ESI 1000fach empfindlicher auf Salzverunreinigungen reagiert als MALDI. Generell lässt sich sagen, dass RNA eine höhere Tendenz zur Kationenanlagerung besitzt als DNA, wobei sich dieser Effekt mit steigender Kettenlänge verstärkt und bei nano-ESI deutlicher auftritt als bei MALDI. Das hat zur Folge, dass bei RNA ein höherer Aufreinigungsgrad notwendig ist, um im Vergleich zu DNA vergleichbare Ergebnisse zu erhalten. Dem entsprechend wurden neben der Aufreinigungsmethode mittels Ionenpaar-HPLC auch verschiedene Pipettenspitzen, die mit Chromatographiematerial gefüllt sind, zur Aufreinigung verwendet. Mit beiden Methoden konnte eine vergleichbare Reduktion der Kationenanlagerungen im Spektrum erreicht werden, wobei klar ist das die Verwendung von Pipettenspitzen die zeit- und kosteneffizientere Lösung darstellt. Durch ein neuartiges Aufreinigungsprotokoll für Pipettenspitzen konnte der Zeitaufwand noch einmal deutlich reduziert werden. Für die Untersuchung nichtkovalenter Oligonukleotid-Komplexe wurden „native“ Bedingungen gefunden, bei der die native Struktur des Komplexes soweit erhalten bleibt, dass ein spezifischer Nachweis mittels nano-ESI möglich ist. Diese Bedingungen stellen einen Kompromiss zwischen einerseits der Stabilität des Komplexes in Lösung und andererseits den optimalen Bedingungen für die Elektrospray-Ionisierung dar. Die erste untersuchte Komplexklasse waren Oligonukleotid-Dimere. Es wurden verschiedenste DNA-, RNA- sowie DNA-RNA-Hetero-Dimere mit Bindungsenergien zwischen 0 und -22,1 kcal/mol mittels nano-ESI vermessen. Es wurde eine klare Grenze ermittelt, bei der die Detektion nichtkovalenter Komplexe möglich ist. Spezifischen Dimere können im Spektrum nur dann detektiert werden, wenn deren Bindungsenergie mindestens -11,5 kcal/mol oder mehr beträgt. Als zweite Gruppe wurden Oligonukleotid-Dimere mit „Minor Groove Binding Ligands“ (MGBLs) mittels nano-ESI untersucht. Es konnte gezeigt werden, dass die MGBLs spezifisch und in der entsprechenden Bindungsstöchiometrie an DNA-Dimere binden. Für einen neusynthetisierten Ligand konnte erstmals die Bindungspräferenz an DNA-Dimere mit der Massenspektrometrie bestätigt werden. In allen Fällen wurde keine unspezifische Bindung an RNA-Dimere oder DNA-RNA-Hetero-Dimere beobachtet. Als letztes wurden RNA-Aminoglykosid-Komplexe mit nano-ESI untersucht. Hierbei konnten nur teilweise spezifischen Komplexe im Spektren nachgewiesen werden, da die Bindungsenergie einiger Komplexe deutlich unterhalb der Nachweisgrenze von -11,5 kcal/mol lag. Alle genannten nichtkovalenten Komplexe wurden auch mittels MALDI vermessen, allerdings konnten keine spezifischen Komplexe nachgewiesen werden. Durch direkten Vergleich von MALDI mit nano-ESI und entsprechende Kontrollexperimente konnte erstmals gezeigt werden, dass die nichtkovalenten Komplexe nicht wie vermutet während der MALDIKristallisation zerstört werden. Viel mehr werden die Komplexe im MALDI-Kristall unzerstört eingebaut und eine Zerstörung erfolgt erst während des Desorptions-/Ionisationsprozesses. Es besteht die berechtigte Vermutung, dass bei stärker bindenden Komplexen eine unzersetzte Ionisation mittels MALDI erfolgen kann. Daher sind weitere Untersuchungen in diese Richtung sinnvoll und es werden zur Zeit Komplexe, deren Bindungsenergie weit über -22,1 kcal/mol liegt, in der Arbeitsgruppe untersucht. Abschließend ist festzustellen, dass die Massenspektrometrie eine schnelle und effiziente Möglichkeit zur Analyse von DNA und RNA ist, die allerdings einen hohen Anspruch an die Sauberkeit und damit an die Aufreinigung der Probe stellt. Darüber hinaus ist die MS auch in der Lage nichtkovalente Oligonukleotid-Ligand-Komplexe zu untersuchen. Da in den letzten Jahren DNA und RNA als mögliches Zielmolekül für neue Therapieansätze mehr und mehr an Bedeutung gewonnen haben, bietet sich die Massenspektrometrie als eine Möglichkeit zur Analyse von Oligonukleotid-Wirkstoff- Komplexe an.
Gepaarte assoziative Magnetstimulation (PAS) kann im primären menschlichen Motorkortex (M1) sowohl langzeitpotenzierungs- (LTP) als auch langzeitdepressionsähnliche (LTD) Erregbarkeitsveränderungen hervorrufen. Dies kann durch die Untersuchung magnetisch evozierter Potentiale (MEP) erfasst werden. Dagegen ist wenig über die Auswirkungen von PAS auf willkürliche Aktivität des motorischen Kortex bekannt. Im ersten Experiment haben wir bewegungsabhängige kortikale Potentiale (MRCP) bei zehn gesunden Probanden im EEG registriert, um die willkürliche Aktivität im Motorkortex während der Vorbereitung zweier motorischer Aufgaben zu erfassen. Die Probanden mussten dabei entweder den Daumen abduzieren (Hauptmuskel: Musculus abductor pollicis brevis, APB) oder das Handgelenk strecken (Hauptmuskel: Musculus extensor carpi radialis, ECR). Die Amplituden der motorisch evozierten Potentiale im APB wurden dabei durch PASLTP gesteigert, durch PASLTD vermindert und blieben bei PAScontrol unverändert. Im Gegensatz dazu wurden sie im ECR durch keine PAS-Bedingungen verändert. PASLTP verminderte die Negativität der MRCP während des späten Bereitschaftspotentials (-500 bis 0 ms vor Bewegungsbeginn) nur in der APB-Aufgabe. Diese Veränderungen zeigten sich hauptsächlich über zentralen Elektroden kontralateral zur bewegten Hand. Dieser Effekt korrelierte negativ mit dem durch PASLTP induzierten MEP-Anstieg im APB. PASLTD und PAScontrol hatten dagegen keinen Einfluss auf die MRCP Amplituden. Unsere Ergebnisse deuten auf eine spezifische Wechselwirkung von PAS mit willkürlicher Aktivität im Motorkortex während der Vorbereitung motorischer Aufgaben hin. Dies könnte durch ein Zusammenspiel aus erhöhter Exzitabilität von M1 und einer unterbrochenen effektiven Konnektivität zwischen prämotorischen Arealen und M1 erklärt werden. Die Modulation des dorsolateralen prämotorischen Kortex (PMd) durch repetitive transkranielle Magnetstimulation (rTMS) verändert die kortikospinale Erregbarkeit in M1. Die Auswirkungen von PMd-rTMS auf vorbereitende Prozesse für willkürliche Bewegungsabläufe sind jedoch unklar. Contingent negative variation (CNV) repräsentiert im EEG kortikale Vorbereitungsprozesse äußerlich getriggerter Bewegungen während das Bereitschaftspotential (BP) Vorbereitungsprozesse intern getriggerter Bewegungen repräsentiert. Im zweiten Experiment wurden CNV und BP jeweils vor und nach PMd-rTMS untersucht. Das Experiment bestand aus drei CNV-Versuchsblöcken mit insgesamt 243 Durchgängen. Dabei mussten die Probanden auf visuelle Anweisung hin eine zwei-Item Finger-Bewegungssequenz durchführen. RTMS wurde sowohl mit 1 Hz als auch mit 5 Hz bei einer Intensität von 110% der aktiven motorischen Schwelle (AMT) unter individueller MR-Navigation appliziert. Die Erfassung des BP erfolgte während der Durchführung derselben motorischen Aufgaben, allerdings bekamen die Probanden keine Anweisungen. Die Durchschnittsamplituden der frühen und späten Komponente von CNV (CNV1:1500-500 ms vor dem Startsignal (S2); CNV: 500-0 ms vor S2) und der frühen und späten Komponente des BP (BP1: 1500-500 ms vor EMG Beginn; BP2: 500-0 ms vor EMG-Beginn) wurden quantitativ für 25 zentrale Elektrodenpositionen verglichen. CNV2 zeigte eine signifikante Bahnung über dem frontal-zentralen Bereich nach 1 Hz PMd-rTMS, blieb aber unverändert nach 5 Hz PMd-rTMS. CNV1, BP1 und BP2 blieben durch 1 Hz und 5 Hz PMd-rTMS unbeeinflusst. Diese Ergebnisse lassen vermuten, dass der dominante PMd eine wichtigere Rolle in der Vorbereitung extern getriggerter Bewegungen spielt, als dies bei intern getriggerten Bewegungen der Fall zu sein scheint. Die CNV2-Antwort könnte eine intensive Interaktion innerhalb des menschlichen motorischen Kontrollnetzwerks anzeigen, die möglicherweise auf kompensationsähnlichen Mechanismen beruht.
The NS5B protein of the hepatitis C virus (HCV) is a RNA-dependent RNA polymerase, which is the key enzyme for viral replication. It is recognized as one of the promising targets for antiviral intervention within the new HCV treatment approach of direct-acting antivirals (DAA). However, several of the known non-nucleoside HCV polymerase inhibitors (NNIs) identified by screening approaches show limitations in the coverage of all six major HCV genotypes (GT). Genotypic profiling therefore has to be implemented early in the screening cascade to discover new broadly active NNIs. This implies knowledge of the specific individual biochemical properties of polymerases from all GTs which is to date limited to GT 1 only. The work submitted here gives a comprehensive overview of the biochemical properties of HCV polymerases derived from all major GTs 1 - 6. Biochemical analysis of polymerases from 38 individual sequences revealed that the optima for monovalent cations, pH and temperature were similar between the GTs, whereas significant differences concerning concentration of the preferred cofactor Mg2+ were identified. Implementing the optimal requirements for the polymerases from each individual GT led to significant improvements in their enzymatic activities. However, the specific activity was distributed unequally across the GTs and could be ranked in the following descending order: 1b, 6a > 2a, 3a, 4a, 5a > 1a. Furthermore, the optimized assay conditions for GT profiling were confirmed by testing the inhibitory activity of four known prototype NNIs, each addressing one of the four NNI binding sites. Additionally, a novel NNI chemotype - identified by screening - is described, the substituted N-phenyl-benzenesulphonamides (SPBS). This inhibitor class showed reversible inhibition of NS5B from HCV 1b Con1 with IC50 values up to 39 nM. Based on the decreased inhibitory activity against a recombinant NS5B protein carrying the mutation L419M, it was assumed that the SPBS inhibitors bound to the thumb site II as it has been described for the carboxy thiophene inhibitors. The postulated binding site was consequently confirmed by analysing a provided co-crystal structure of NS5B in complex with a SPBS analogue. Notably, the two SPBS analogues SPBS-1 and SPBS-2 reported here revealed significant differences in addressing the NH-group of the main chain Y477 by hydrogen-bonds, watermediated or directly, which provoked a shift of the carboxyphenyl group of the inhibitors towards the H475 position for the water-mediated binding mode. Interestingly, the differences observed in the binding mode led to a different cross resistance profile at positions M423 and I482. Using the previously optimized biochemical primer-dependent transcription assay, inhibitory activity of the SPBS could be demonstrated against polymerases from HCV GTs 1a and 1b whereas the inhibitor class failed to inhibit any of the non-GT 1 polymerases. Furthermore, initial antiviral activity for SPBS was demonstrated against the subgenomic replicons of HCV GTs 1a and 1b, respectively, and no considerable cytotoxic potential against a panel of ten different cell types. Finally, concerning a possible future treatment without PEG-IFN α or ribavirin, the SPBS analogues were found to display additive to synergistic effects in combination with the benzothiadiazine, the benzofuran and the indole - representative inhibitors for the binding sites palm I, palm II and thumb I, repectively - in the biochemical assay. Within the same binding site as the SPBS, the reference compound hydroxydihydropyranone displayed additive interactions only with the benzothiadiazine (palm I) in the biochemical assay as well as in cell culture. Hence it could be concluded that, having characterized one individual NNI, no universal predication is possible concerning the combinatory behaviour of NNIs binding to the same binding site. As synergistic, antagonistic or additive interactions are inhibitor-dependent (not binding sitedependent) each novel NNI has to be characterized individually in one-to-one combinations.
The long sought molecular function of membrane raft-associated flotillin proteins is slowly becoming resolved, partially owing to the increasing knowledge about their interaction partners. Being ubiquitously expressed and evolutionarily highly conserved, flotillins carry out important cellular functions, one of which is the regulation of signal transduction pathways. This study shows that the signaling adaptor protein fibroblast growth factor receptor substrate 2 (FRS2) directly interacts both in vivo and in vitro with flotillin-1 (flot-1). FRS2 is an important docking protein of many receptor tyrosine kinases. It regulates downstream signaling by forming molecular complexes with other adaptor proteins and tyrosine phosphatases, and seems to be a critical mediator of sustained extracellular signal regulated kinase (ERK) activity. Flot-1 has also been implicated in the regulation of ERK activity upon EGF and FGF stimuli. Furthermore, flot-1 forms signalosomes with EGFR and the downstream components of the MAP kinase pathway. The newly discovered interaction between FRS2 and flot-1 was shown to be mediated by the phosphotyrosine binding (PTB) domain and, to a lesser extent, the C-terminus (CT) of FRS2 and by the C-terminus of flot-1. Flot-1 coprecipitated together with FRS2 from murine tissues and cell lysates, demonstrating that this interaction also takes place in vivo. Interestingly, flot-2, which shows a high homology to flot-1 and forms stable oligomeric complexes with it, does not appear to directly interact with FRS2. Novel insights into the functional role of the interaction between flot-1 and FRS2 were provided by the results showing that depletion of flot-1 affects the cellular localization of FRS2. In hepatocytes stably depleted of flot-1, FRS2 appeared to be more soluble. Furthermore, upon pervanadate stimulation of the cells, a small fraction of FRS2 was recruited into detergent resistant membranes, but the recruitment did not take place in the absence of flot-1. Triggered by the same stimulus, a fraction of FRS2 was translocated to the nucleus independently of flot-1. Overexpression of FRS2 has previously been shown to result in increased ERK activation. However, in cells depleted of flot-1, FRS2 was not able to compensate for the compromised ERK activation after EGF or FGF stimulation. This might imply that FRS2 and flot-1 are functionally interconnected and that FRS2 resides upstream of flot-1. Taken together, the results presented here indicate that this complex may be involved in the control of signaling downstream of receptor tyrosine kinases and is important for ensuring a proper signaling response. In the absence of flot-1, increased Tyr phosphorylation of FRS2 was observed. It is known that Tyr and Thr phosphorylation of FRS2 are reciprocally regulated. Since ERK is a known executor of the FRS2 Thr phosphorylation, and ERK activity was shown to be severely diminished upon flot-1 depletion, the increased Tyr phosphorylation of FRS2 was in agreement with this and might be a direct consequence of a decreased ERK activity upon flot-1 depletion. FRS2 owes its name to the major and the first described function of this protein as a substrate for FGFR. PTB domain of FRS2 was published to constitutively bind the juxtamembrane domain of FGFR. In this study, the PTB domain was mapped to be involved in the constitutive interaction with flot-1 and the competition was shown to exist between flot-1 and FGFR1 for binding to FRS2. Another novel interaction partner of FRS2 was discovered in the present study. Cbl-associated protein (CAP) is an adaptor protein with three SH3 domains and it plays a role during insulin signaling by recruiting the signaling complex to lipid rafts. CAP was previously shown to interact with flot-1 via the SoHo domain, and this interaction was found to be crucial for the lipid raft recruitment of other signaling components. Both the PTB domain and CT of FRS2 were found to mediate the interaction with CAP, whereas in CAP, the SoHo domain, together with the third SH3 domain, seems to bind to FRS2. SH3 domains mediate the assembly of specific protein complexes by binding to proline rich sequences, several of which are present in FRS2. Due to overlapping interaction domains, FRS2 and flot-1 competed for the binding to CAP. However, the interaction with neither CAP nor flot-1 was necessary for the observed nuclear translocation of FRS2. Since CAP is expressed as several tissue- and developmental stage-specific isoforms, a further aim of this study was to analyze the expression of its isoforms in mouse embryonic fibroblasts (MEFs). Many new isoforms were discovered here which have not been described in the literature so far. They all contain the SoHo domain and three SH3 domains, but differ among themselves by the presence and length of a proline-rich region that preceeds the SoHo domain and by a novel 20-amino acid (AA) stretch between the second and the third SH3 domain. The length of the proline-rich region turned out to be an important factor determining the strength of the interaction with FRS2. The interaction was found to be weakened by the increasing length of this region. The new isoforms possessing the 20-AA stretch are specifically expressed in murine muscular tissues, with the highest level in the heart. During adipogenesis, we observed a shift in the abundance of the isoforms, in that only the isoforms without the insertion were shown to be upregulated on mRNA level. However, during myogenesis, preferentially expressed isoforms were those with the insertion. The collected data implicate that isoforms with the 20-AA insertion might be more ubiquitous in nondifferentiated/embryonic cells and that the observed "isoform-switch" might be dependent on the cell fate and differentiation state.
Molekulare Regulation der UDP-Zucker-Biosynthese Untersuchungen anhand der myo-Inositoxygenase

Der Nukleotidzucker UDP-Glucuronsäure ist die prinzipielle Zuckervorstufe für UDP-Galacturonsäure, -Xylose, -Arabinose und –Apiose, welche alle in die Zellwandpolymere der pflanzlichen Zellwand einfließen. UDP-Glucuronsäure kann in Arabidopsis über zwei alternative, funktionelle Synthesewege gebildet werden, wobei entweder die UDP-Glucose-Dehydrogenase oder die myo-Inositoxygenase als Schlüsselenzym, welche den Kohlehydratfluss in Richtung Zellwandbiosynthese katalysiert, involviert ist. In dieser Arbeit wurden die Gene für die Enzymisoformen der Inositoxygenase (MIOX) aus Arabidopsis thaliana analysiert. Sie repräsentieren eine kleine Genfamilie bestehend aus vier Isoformen. Studien mittels Promotor::GUS-Konstrukten und RT-PCR zeigten, dass die Transkription der MIOX-Gene eine sehr transiente und organspezifische Genexpression ist. Die Isoformen MIOX1 und MIOX2 ließen sich in nahezu allen Geweben nachweisen wogegen die Isoformen MIOX4/5 nur in den generativen Geweben aufzufinden waren. Es konnte eine deutliche Präsenz aller vier MIOX-Isoformen in den generativen Geweben nachgewiesen werden und die Nukleotidzucker, die während der Samenentwicklung benötigt werden, scheinen überwiegend über die MIOX zur Verfügung gestellt zu werden. So zeigen T-DNA-Insertionslinien von ΔMIOX1 & 2 eine reduzierte Schleimhülle um die Samen und eine erhöhte Ausfallrate bei der Samenentwicklung auf. Ansonsten zeigen die untersuchten T-DNA-Insertionslinien einen ähnlichen Wuchs wie der Wildtyp auf. Auch konnten keine Unterschiede über die durchgeführten Zellwandanalysen, mittels GC-MS, MALDI und Dionex-HPLC verifiziert werden, was durch die Redundanz der MIOX- und UGD-Isoformen erklärt werden könnte. Nichtsdestotrotz konnte bei den Isoformen ΔMIOX1 & 2 eine dramatische Reduktion beim Einbau von 3H-Inosit in Zellwandpolymere von Keimlingen verzeichnet werden, was einen klaren Beweis für eine funktionelle MIOX liefert, da in diesem Gewebe diese beiden Isoformen die einzig aktiven sind. Außerdem konnten über Promotor-Deletions-Analysen potentielle cis-Elemente für die Promotoren von MIOX2 und MIOX4 aufgedeckt werden.
Die Verbreitung westlicher Verhaltens- und Lebensmuster im Rahmen der Globalisierung führt zu einer dramatischen Zunahme des Typ 2 Diabetes. Eine schwerwiegende Komplikation der diabetischen Erkrankung ist die Wundheilungsstörung, deren molekulare Pathophysiologie noch weitgehend unverstanden ist. Voraussetzung für einen koordinierten Wundheilungsprozess ist die ausgewogene Interaktion wundheilungsrelevanter Faktoren, die eine Vielfalt an Signaltransduktionskaskaden induzieren und somit zu einem streng kontrollierten Heilungsprozess beitragen. Darüber hinaus scheint auch die Insulinsensitivität der Haut für den Heilungsverlauf eine essentielle Rolle zu spielen. Da die Proteinkinase B/Akt nicht nur ein zentrales Molekül der Insulin-Signaltransduktion ist, sondern als Knotenpunkt vieler Signalkaskaden im Mittelpunkt zellulärer Ereignisse steht, sollte in der vorliegenden Arbeit die Rolle und Funktion der Proteinkinase Akt in der kutanen Wundheilung untersucht werden. Sowohl in der Haut als auch im Wundgewebe konnte Akt1 als dominante Isoform identifiziert werden. Die Verletzung des Hautgewebes induzierte einen Anstieg der Akt1-Expression und -Phosphorylierung in der Epidermis akut heilender Wunden. Insbesondere die am Wundrand gelegenen Keratinozyten waren durch eine starke Expression der Akt1-Kinase gekennzeichnet. Die Phosphorylierung und somit Aktivierung der Akt1-Kinase stieg im Verlauf der Wundheilung stetig an und erreichte ihr Maximum in der Endphase des Heilungsprozesses. Begleitet wurde die Aktivierung dieser Kinase von einer Phosphorylierung des eIF4E-BP1 und einer starken VEGF-Sekretion. Dagegen konnte in diabetisch chronischem Wundgewebe weder eine Aktivierung der Akt1-Kinase noch eine Phosphorylierung des eIF4E-BP1 nachgewiesen werden, was mit einer deutlich verminderten VEGF-Sekretion einherging. Um nun zu untersuchen, ob im Prozess der Wundheilung die VEGF-Sekretion in einem funktionellen Zusammenhang zur Akt1-Aktivierung steht, wurde in vitro der Einfluss wundheilungsrelevanter Faktoren, wie EGF, einer Kombination proentzündlicher Zytokine oder Insulin auf die Aktivierung der Akt1-Kinase und VEGF-Biosynthese untersucht. Obwohl alle Faktoren sowohl eine Aktivierung der Akt1-Kinase als auch VEGF-Sekretion induzierten, wurde ausschließlich die Insulininduzierte VEGF-Biosynthese über den PI3-Kinase/Akt-Signalweg vermittelt. Die Regulation der Insulin-induzierten VEGF-Biosynthese erfolgte posttranskriptionell aus einem gleichbleibenden Pool an VEGF-mRNA über die Phosphorylierung des eIF4E-BP1. Durch die Überexpression einer konstitutiv aktiven Akt1-Mutante und die Verwendung des mTOR-Inhibitors Rapamycin konnte mTOR als Mediator der Akt1-vermittelten Phosphorylierung des eIF4E-BP1 und somit der VEGF-Biosynthese identifiziert werden. In vitro-Lokalisationsexperimente zeigten, dass eine vollständig phosphorylierte und somit aktive Akt1-Kinase nach Insulinstimulation im Zytoplasma lokalisiert ist - exakt dort, wo die Regulation der Translation stattfindet. Zusammenfassend weisen diese Ergebnisse darauf hin, dass Insulin im kutanen Heilungsverlauf die VEGF-Biosynthese posttranskriptionell über eine Akt1-vermittelte Phosphorylierung des eIF4E-BP1 induzieren kann. Eine ausbleibende Akt-Aktivierung in insulinresistenten Keratinozyten könnte somit zu einer verminderten VEGF Sekretion und folglich zu einer verzögerten Angiogenese in chronisch diabetischen Wunden beitragen.
Da zwischen der bakteriellen und der archaeellen Selenoprotein-Biosynthese deutliche Unterschiede existieren, sollten in dieser Arbeit trans-aktive Faktoren der Selenoprotein-Biosynthese von M. maripaludis molekulargenetisch analysiert werden um weitere Einblicke in die archaeelle Selenocystein-Biosynthesestrategie zu gewinnen. Die folgenden Ergebnisse wurden durch diese Arbeit erhalten: In Archaeen muss der Selen-Donor wie in Bakterien zu Selenophosphat phosphoryliert werden. Ohne die Aktivierung des Selen-Donors sind Archaeen nicht in der Lage Selenoproteine zu synthetisieren. Das für die Phosphorylierung des Selen-Donors verantwortliche Protein stellt in M. maripaludis interessanterweise selbst ein Selenoprotein dar. M. maripaludis ist nicht zwingend auf die Synthese von Selenocystein in einer Zwei-Schritt-Methode ausgehend von Seryl-tRNAsec, durch die Enzyme PSTK und SepSecS, angewiesen. Das Einbringen des bakteriellen Enzyms Selenocystein-Synthase in M. maripaludis, welches die gleiche Reaktion in einem Schritt durchführt, ohne Bildung des Intermediats O-Phosphoseryl-tRNAsec, ermöglicht es dem Organismus ebenfalls Selenoproteine zu bilden. Dies zeigt, dass das Intermediat O-Phosphoseryl-tRNAsec der archaeellen Selenoprotein-Biosynthese-Strategie für M. maripaludis nicht essentiell ist. Durch die Disruption eines notwendigen Faktors in der Selenoprotein-Biosynthese wird in M. maripaludis eine Veränderung der mRNA-Mengen, der für Selenoproteine und deren Cystein-haltigen Isoformen codierenden Gene ausgelöst. Diese Regulation ist unter bestimmten Bedingungen nicht reversibel. Ein Unterschied hinsichtlich der Essentialität des selenierenden Systems konnte zwischen den Stämmen M. maripaludis JJ und M. maripaludis S2 festgestellt werden. In M. maripaludis S2 ist das selenierende System essentiell. M. maripaludis JJ hingegen ist nicht auf Selenoproteine angewiesen. Weder die physiologische noch die molekulare Grundlage konnten im Rahmen dieser Arbeit aufgeklärt werden, allerdings kann das neu identifizierte HesB-ähnliche Selenoprotein als verursachender Faktor ausgeschlossen werden.
In der vorgelegten Arbeit wurden 50 Mutationen von Aminosäuren im Bereich der Chinoloxidationsbindungsstelle (Qo-Site) des bc1-Komplexes aus Paracoccus denitrificans untersucht. Hierzu wurden die Mutanten erstellt, Kinetiken der Substratbindung bestimmt und EPR Spektren aufgenommen. Mit den gleichen Methoden wurden auch verschiedene Klasse I und Klasse II Inhibitoren der Qo-Site untersucht. Wenngleich der Reaktionsmechanismus auch mit Hilfe dieser Mutationen nicht vollständig aufgeklärt werden konnte, so konnten doch neue Einsichten in die Funktionsweise des bc1-Komplexes gewonnen werden. Verschiedene Modelle wurden vorgeschlagen, um die energetisch günstige aber thermodynamisch unwahrscheinliche Verzweigung der beiden Elektronen des Ubichinols auf die beiden Redoxketten des bc1-Komplexes der Atmungskette zu beschreiben. Da die Redoxpotentiale der beiden primären Elektronenakzeptoren extrem unterschiedlich sind, scheint es zwingend zu sein, dass beide Elektronen den Hochpotentialzweig (Rieske, Cytochrom c1, Cytochrom c) durchlaufen. Energetisch bietet jedoch die Wiederverwendung eines der beiden Elektronen den immensen Vorteil der realen Verdopplung der Protonenpumpleistung. Die wichtigsten Mutationen der vorliegenden Arbeit betreffen die vermuteten direkten Bindungspartner des Ubichinols, bE295 und fH155. Das Histidin ist Teil der beweglichen Rieske Untereinheit des bc1-Komplexes, das Glutamat ist Teil eines hoch konservierten Abschnitts des Cytochrom b. Aufgrund der gemessenen Wirkungen der untersuchten Mutationen auf die kinetischen Kennzahlen und EPR Parameter der eingesetzten Substrate und Inhibitoren lassen sich folgende Schlüsse ziehen. Die Bindung des Chinol-Substrats in der Qo-Site benötigt die korrekte Ausrichtung und den unveränderten Bindungspartner fH155 der Rieske [2Fe-2S]-Gruppe. Das Fehlen des zweiten vermuteten Bindungspartners bE295, der ein Teil des hochkonservierten PEWY-Loops ist, wird hingegen toleriert. Die Aktivitäten bei Mutationen dieser Aminosäure liegen mit bis zu 56% im Vergleich zum Wildtyp (bE295A) relativ hoch. Auch unter Berücksichtigung von möglichen bypass Reaktionen ist es fraglich, ob das Glutamat des PEWY-Loops ein direkter Bindungspartner des Substrats ist. Der Inhibitor Stigmatellin bindet im bc1-Komplex von Paracoccus denitrificans irreversibel in der Qo-Site, auch wenn einer der beiden vermuteten Bindungspartner (bE295 bzw. fH155) nicht verfügbar oder verändert ist. Daraus kann man schließen, dass die Bindung des Stigmatellins im Wesentlichen von seiner Seitenkette im Bereich des Chinol-Eintrittskanals des bc1-Komplexes stabilisiert wird. Wechselwirkungen der bizyklischen Kopfgruppe mit Aminosäuren der Chinoloxidationsbindungstasche sind hierfür nicht zwingend notwendig. Weitere durchgeführte Mutationen lassen folgende weitere Rückschlüsse zu: Der korrekte Bindungsraum der Qo-Site ist für eine maximale Umsatzgeschwindigkeit des bc1-Komplexes erforderlich. Dies zeigen Mutationen von bG158 und bV161. Die Struktur des hochkonservierten PEWY-Loops ist mitentscheidend für die Aktivität des bc1-Komplexes. Störungen der Konfiguration in diesem Bereich führen zu einer deutlichen Abnahme der Aktivität. Dies lassen Mutationen von bY297, bP294 und bW296 erkennen. Der ef-Loop hat einen Einfluss auf die Bewegungsvorgänge der Rieske-Kopfgruppe. Er ist jedoch nicht sehr flexibel (bL286H). Veränderungen im Wasserstoffbrückennetzwerk der Rieske-Kopfgruppe führen zu einer deutlichen Verminderung der Elektronentransferraten (bY159F, bS157A und bF151H). Die Bindungsregionen von Klasse I Inhibitoren in der Qo-Site überlappen mit denen des natürlichen Substrats. Sie sind jedoch nicht identisch, was sich im unterschiedlichen Verhalten einiger Mutationen auf die Aktivität von Substrat und die Inhibitionswirkung von Klasse I Hemmstoffen ausdrückt. Die Untersuchungen der Auswirkungen von Mutationen auf einen vermuteten Wasserkanal im Bereich des Häm bL und auf eine angenommene Zinkbindungsstelle im gleichen Bereich zeigen keine eindeutig interpretierbaren Ergebnisse. Die Mutationsstudien im Bereich des Cytochrom c1 wurden begonnen. Zusätzliche Mutanten in diesem Bereich könnten weiterführende Aufschlüsse über die Wechselwirkung der Rieske-Kopfgruppe mit Cytochrom c1 bringen. Die vorgelegte Studie lässt erkennen, dass die Vorgänge in der Qo-Site des bc1-Komplexes aus Paracoccus denitrificans höchst komplex sind. Zusätzliche Untersuchungen der erzeugten Mutanten mit Hilfe weiterer Detektionsmethoden, insbesondere CD- und FTIR-Messungen, könnten in Zukunft eine noch bessere Einsicht in die Vorgänge dieses wichtigen Komplexes der Atmungskette geben.
Teerfleckenpilze (Ascomycota, Phyllachorales) kommen weltweit vor, haben aber einen deutlichen Verbreitungsschwerpunkt in den Tropen. In dieser Studie wird die Diversität der Phyllachorales in Panama untersucht und taxonomische Grundlagenforschung durch molekulare Untersuchungen und ökologische Beobachtungen ergänzt. Arten der Phyllachorales bilden schwarz glänzende Flecken oder Krusten auf Blättern und Stängeln von Pflanzen. Die Fruchtkörper dieser Mikropilze sind meistens in das Wirtsgewebe eingesenkt, können aber auch oberflächlich angelegt sein. Die Größe der teerfleckenähnlichen Infektionen variiert zwischen 0,2 mm bis zu mehreren Zentimetern. Teerfleckenpilze mit eingesenkten Fruchtkörpern entwickeln in den meisten Fällen schildförmige, epidermale Deckstrukturen oberund/oder unterhalb der Perithecien. Die unitunicaten Asci können einen kleinen apikalen Ring besitzen, der sich in Jodlösung nicht blau färbt. Die hyaline Ascosporen sind meistens glatt und können von einer schleimigen Hülle umgeben sein. Die assoziierten Andromorphstadien sind durch fadenförmige Spermatien gekennzeichnet. Nur wenige morphologische Merkmale werden als verlässlich betrachtet und stehen für die Bestimmung von Arten zur Verfügung. Derzeit sind 1.226 Arten von Phyllachorales weltweit beschrieben. Puerto Rico gilt als eines der am besten untersuchten Länder in Bezug auf Teerfleckenpilze. Ungefähr 100 Arten sind derzeit von Puerto Rico bekannt, weitere Erstnachweise werden erwartet. Im Gegensatz dazu wurden in Panama bisher nur wenige mykologische Untersuchungen durchgeführt. Zwischen 1927 und 1991 wurden nur 39 Teerfleckenpilze nachgewiesen, obwohl die Landfläche Panamas achtmal größer ist als die von Puerto Rico und eine fast viermal höhere Pflanzendiversität aufweist. Aufgrund der Vielzahl potentieller neuer Wirtspflanzen in Panama und des von Teerfleckenpilzen bevorzugten tropischen Klimas wird eine weitaus höhere Anzahl an Arten der Phyllachorales erwartet. Zwischen 2005 und 2008 wurden mehreren Exkursionen in den Provinzen Bocas del Toro und Chiriquí durchgeführt, um die tatsächliche Diversität pflanzenparasitischer Mikropilze in Panama zu untersuchen. Über 1.000 Belege wurden von insgesamt vier Wissenschaftlern während 7 Sammelreisen von jeweils 2‐4 Wochen gesammelt. 185 Belege zeigen mehr oder weniger deutliche Symptome von Teerfleckenkrankheit und entsprechen 42 Arten der Phyllachorales, die bisher noch nicht für Panama bekannt sind. 81 Teerfleckenpilze aus Panama werden in dieser Arbeit mit detaillierten Beschreibungen, Zeichnungen und Fotos vorgestellt. Von den 66 bis zur Art bestimmten Phyllachorales werden 27 erstmalig für Panama, 19 für Zentralamerika und 3 für die Neue Welt genannt. Die Erstnachweise von Camarotella costaricensis, Coccodiella miconiicola, Ophiodothella galophila, Phyllachora amphibola, Ph. engleri und Ph. zanthoxylicola wurden bereits vorab publiziert. 21 Arten werden hier zum ersten Mal vorgestellt: Catacauma paramoense, Coccodiella miconiae, Ophiodothella cuervoi, Phyllachora acaciae subsp. pusaethae, Ph. acalyphae, Ph. araliarum, Ph. balansae, Ph. buddleiae, Ph. cynodontis, Ph. galavisii, Ph. gouaniae, Ph. leeae, Ph. meliosmae, Ph. microtheles, Ph. ocoteae, Ph. paraguaya, Ph. phyllanthophila var. phyllanthophila, Ph. puncta subsp. dalbergiicola, Ph. ruelliae, Ph. serjaniae und Ph. smilacicola. Zusätzlich konnte erstmals der Hyperparasit Perizomella inquinans auf Ph. ocoteae für Panama und der Teerfleckenpilz Ph. guazumae für Costa Rica nachgewiesen werden. Von den 15 bisher noch unbestimmten Teerfleckenpilzen sind 9 Arten der Gattungen Camarotella und Phyllachora wahrscheinlich neu für die Wissenschaft und 6 Arten müssen bis zur Bestimmung noch weiter untersucht werden. 10 Taxa wurden aufgrund von Synonymisierung, Zitierungsfehlern oder mangelhaftem Material ausgeschlossen. Für eine sichere Bestimmung der Arten aus Panama wurden alle weltweit bekannten Teerfleckenpilze auf nah verwandten Wirtspflanzen miteinander verglichen. Dazu wurden intensive Literaturrecherche und detaillierte lichtmikroskopische Studien durchgeführt. 190 Typen und autoritative Belege wurden zusätzlich aus 22 Herbarien weltweit ausgeliehen und untersucht. Dabei wurden einige Gruppen von Teerfleckenpilzen auf bestimmten Wirtsfamilien teilweise überarbeitet und Schlüssel für die Bestimmung der jeweiligen Arten erstellt. Ein Vergleich der charakteristischen Merkmale wird als tabellarische Übersicht in den Anmerkungen vorgestellt. Aufgrund der intensiven taxonomischen Arbeit konnten 7 neue Synonyme aufgedeckt werden. Catacauma contractum ist ein Synonym von Ph. gouaniae, Ph. clypeata von Ph. paraguaya, Ph. insueta von Ph. serjaniae, Ph. paulliniae von Ph. galavisii und Ph. swieteniae von Ph. balansae. Für Ph. roureae werden gleich zwei neue Synonyme vorgeschlagen, Ph. connari und Ph. panamensis. Einige der untersuchten Herbarbelege waren nur bruchstückhaft vorhanden oder in sehr schlechtem Zustand. Viele Belege konnten aufgrund mangelnder Informationen nicht gefunden oder wegen sehr langen Lieferungszeiten oder fehlender Kooperationen noch nicht untersucht werden. Die Erstbeschreibungen vieler Arten in der Literatur sind lückenhaft und für die wenigsten Arten sind Abbildungen vorhanden. Dadurch ist eine sichere Artbestimmung ohne Untersuchung des zugehörigen Herbarmaterials oft unmöglich. In dieser Arbeit werden alle 81 untersuchten Teerfleckenpilze mit detaillierten Beschreibungen, Zeichnungen und Fotografien dargestellt, um zukünftige Bestimmungsarbeiten zu erleichtern. 16 Teerfleckenpilze wurden im Rahmen dieser Arbeit zum ersten Mal illustriert: Catacauma paramoense, Ophiodothella cuervoi, O. galophila, Phyllachora acaciae var. enterolobii, Ph. acalyphae, Ph. araliarum, Ph. bonariensis, Ph. engleri, Ph. galavisii, Ph. leeae, Ph. meliosmae, Ph. ocoteae, Ph. ruelliae, Ph. smilacicola, Ph. verbesinae und Polystigma pusillum. Darüber hinaus wurden Ph. acaciae subsp. pusaethae, Ph. cecropiae, Ph. leptochloae, Ph. puncta subsp. dalbergiicola und Ph. weirii in ihrer Darstellung vervollständigt. Für einige Arten werden bislang unveröffentlichte morphologische Merkmale vorgestellt, z.B. chondroide Hyphen, Schleimhüllen oder Andromorphstadien, und die mögliche Verwendung dieser Merkmale zur Artabgrenzung diskutiert. Zusätzlich zu den morphologischen Merkmalen werden molekulare Daten hinzugezogen. Zu Beginn dieser Arbeit standen Sequenzen von nur acht verschiedenen Arten der Phyllachorales s.str. zur Verfügung, davon 3 aus der Gattung Phyllachora. Aufgrund der Tatsache, dass Arten der Phyllachorales auf lebendes Pflanzenmaterial angewiesen sind, ist es bisher noch nicht gelungen, diese Pilze in Kultur zu halten. Die Isolation von DNA‐Material aus eigenen Herbarbelegen oder von auf Silicagel getrockneten Proben erwies sich als wenig erfolgreich. Es war notwendig, das frisch gesammelte Material direkt vor Ort in Panama zu verarbeiten. Die Extraktion von reinem und geeignetem Pilzgewebe stellte sich aufgrund der sehr geringen Fruchtkörpergröße und der engen Anhaftung an das Pflanzengewebe als sehr mühsam dar. Trotzdem wurden 10 neue DNA‐Sequenzen, die jeweils für die kleine bzw. große ribosomale Untereinheit kodieren, von 7 Teerfleckenpilzen, Coccodiella miconiae, Coccodiella sp., Phyllachora engleri, Ph. graminis, Ph. leeae, Ph. ulei und Ph. zanthoxylicola, auf 10 verschiedenen Wirtspflanzen erfolgreich isoliert. Erste molekulare Untersuchungen mittels Maximum Likelihood, Maximum Parsimony und Bayesianischer Analyse eines kombinierten Datensatzes unterstützen die Zugehörigkeit der Phyllachorales zur Unterklasse Sordariomycetidae und deuten auf eine paraphyletische Entwicklung der Gattung Phyllachora hin. Die Typusart, Phyllachora graminis, könnte mit Arten der Gattung Coccodiella enger verwandt sein als mit anderen Arten der Gattung Phyllachora. Bisher fehlen aber noch entsprechende morphologische Merkmale die diese These unterstützen. Weitere Untersuchungen mit einem erweiterten Datensatz sollten unternommen werden. Aufgrund der biotrophen Lebensweise und der damit verbundenen engen Anpassung an die jeweilige Wirtspflanze wird eine hohe Spezifität der Teerfleckenpilze angenommen. Die Identifikation der Wirtspflanze bildet daher eine wichtige Grundlage bei der Bestimmung des Pilzes. Durch die Abwesenheit fertiler Strukturen, wie Blüten und Früchte, wurde eine eindeutige Bestimmung oft erschwert. Pflanzenspezialisten, Fachliteratur, Herbarmaterial und Internetdatenbanken wurden hinzugezogen, um eine möglichst genaue Bestimmung der Wirtspflanzen zu erreichen. Die 81 für Panama vorgestellten Teerfleckenpilze parasitierten auf ungefähr 93 verschiedenen Wirten aus 72 Gattungen und 43 Pflanzenfamilien. 73 Wirtspflanzen wurden bis zur Art bestimmt, 20 können mit Gattungsnamen angesprochen werden und sind von den bisher bestimmten Arten sicher verschieden. 5 Arten sind derzeit noch gänzlich unbestimmt. Von den 73 vollständig bestimmten Pflanzenarten werden 29 zum ersten Mal als Wirte für den entsprechenden Parasiten vorgestellt: Acalypha diversifolia für Phyllachora acalyphae, Anthurium concinnatum für Phyllachora engleri, Buddleja nitida für Phyllachora buddleiae, Cissus trianae für Phyllachora leeae, Croton draco und Croton hirtus für Phyllachora tragiae, Dalbergia brownei für Phyllachora puncta subsp. dalbergiicola, Dichanthelium acuminatum und Dichanthelium viscidellum für Phyllachora bonariensis Speg., Dicliptera iopus für Phyllachora ruelliae, Dioscorea trifida und Dioscorea urophylla für Phyllachora ulei, Entada polystachya für Phyllachora acaciae subsp. pusaethae, Inga punctata und Inga sierrae für Phyllachora amphibola, Luehea seemannii für Phyllachora paraguaya, Ocotea veraguensis für Phyllachora ocoteae, Oreopanax xalapensis für Phyllachora araliarum, Ossaea micrantha für Coccodiella miconiicola, Paspalum paniculatum und P. pilosum für Phyllachora paspalicola, Paullinia bracteosa für Phyllachora galavisii, Phyllanthus anisolobus für Phyllachora phyllanthophila var. phyllanthophila, Serjania atrolineata für Phyllachora serjaniicola, Serjania mexicana für Phyllachora serjaniae, Vaccinium floribundum für Catacauma paramoense und Ophiodothella cuervoi, Xylosma flexuosa für Trabutia xylosmae und Zanthoxylum melanostictum für Phyllachora zanthoxylicola. Teilweise wurden diese Erstnachweise schon vorab publiziert. Arten der Familien Fabaceae, Melastomataceae und Poaceae sind besonders häufig mit Teerfleckenpilzen infiziert und scheinen bevorzugte Wirtspflanzen für Arten der Phyllachorales zu sein. Für ökologische Studien wurden bestimmte Teerfleckenpilze an ausgesuchten Standorten wiederholt zu verschiedenen Jahreszeiten gesammelt und untersucht. Dabei zeigte sich, dass Arten der Ordnung Phyllachorales in zwei Habitaten besonders häufig anzutreffen sind: (1) in offenen, gestörten Gebieten mit Ruderalvegetation und deutlicher Trockenperiode sowie (2) in dichten, ungestörten Bergregenwäldern mit hoher relativer Luftfeuchtigkeit und kontinuierlichem Niederschlag. Eine hohe Pflanzendiversität korreliert nicht mit einer hohen Diversität an Teerfleckenpilzen. Vergleichende Untersuchungen zeigten, dass das Vorkommen von Arten der Phyllachorales wahrscheinlich stärker vom Vorkommen und der Dichte bevorzugter Wirtspflanzen abhängt als von ökologischen Faktoren wie z.B. Vegetationstyp, Höhenlage, relativer Lichtintensität, durchschnittlichem Jahresniederschlag oder jahreszeitlicher Temperaturentwicklung. Die in dieser Studie vorgestellten Teerfleckenpilze stammen aus nur 5 von 12 Provinzen Panamas: Bocas del Toro und Chiriquí im Westen sowie Colón, Kuna Yala und Panamá im Zentrum des Landes. Mithilfe einer Datenbank wurde eine Vielzahl verfügbarer Literaturdaten mit Internet‐Datensätzen ergänzt und daraus eine Verbreitungskarte von Arten der Phyllachorales in der Welt erstellt. Für viele Länder sind nur wenige oder gar keine Teerfleckenpilze bekannt. Das lückenhafte Vorkommen in Panama und anderen Ländern der Welt entspricht aber keinesfalls der natürlichen Verbreitung dieser Pilze, sondern gibt die unterschiedliche Intensität von Forschungsaktivitäten in Bezug auf die Teerfleckenpilze wieder. Weitere Erstnachweise und neue Arten der Ordnung Phyllachorales werden für Panama und die Welt erwartet, denn unser Wissenstand über diese Gruppe ist sehr lückenhaft und weite Gebiete sind bislang noch unzureichend untersucht.
Studies in particular of the last decade showed that active neurogenesis continuously takes place in the subventricular zone (SVZ) of the lateral ventricles of the adult rodent brain. Neurogenesis in the SVZ leads to migration of neuroblasts within the rostral migratory stream (RMS) and mature neuron formation mainly in the olfactory bulb (OB). According to present understanding, glial cells with astrocytic properties represent the actual adult neural stem cells. The cell types representing the various cellular transition states leading to the formation of mature neurons as well as the mechanisms controlling adult neurogenesis and neuroblast migration are poorly understood. A previous study from this laboratory demonstrated that the ATP-hydrolyzing enzyme nucleoside triphosphate diphosphohydrolase 2 (NTPDase2) is associated with type B cells, the presumptive neural stem cells. NTPDase2 is a protein of the plasma membrane with its catalytic site facing the extracellular space. It hydrolyzes extracellular nucleoside triphosphates to their respective nucleoside diphosphates. This raises the possibility that the signaling pathway via extracellular nucleotides is involved in the control of adult neurogenesis. Neurons as well as glial cells express several subtypes of receptors (P2 receptors) that are responsive to the nucleotides ATP, ADP, UTP, or UDP. P2X receptors are ATP-gated Na+, K+ and Ca2+ permeable ion channels, P2Y receptors are coupled to trimeric G-proteins. In order to probe for a functional role of nucleotides in adult neurogenesis, the present study referred to an in vitro system (neurospheres). Neurospheres produced from isolates of the mouse SVZ and cultured in the presence of EGF and bFGF expressed the neural stem cell marker nestin and also GFAP, S100β, NTPDase2 and tissue non-specific alkaline phosphatase. Neurospheres generated from the cells of the subventricular zone were multipotenital. This was revealed by immunostaining of differentiated cells with markers for astrocytes, neurons and oligodendrocytes. The presence of ecto-nucleotidase was verified by analyzing the free phosphate released from nucleotides. The tissue non-specific form of alkaline phosphatase was the predominant enzyme. Both NTPDase2 and TNAP could be identified by immunocytochemistry and Western blotting. Hydrolysis was not observed for p-nitrophenyl thymidine monophosphate, a substrate of members of the ectonucleotide pyrophosphatase/phosphodiesterase family (NPP1 to NPP3). Since ecto-nucleotidases control the availability of extracellular nucleotide agonists, neurospheres were studied for the potential expression and functional role of nucleotide receptors. Neurospheres responded to extracellular nucleotides with a transient rise in Ca2+ (ATP = ADP > UTP). The rise in Ca2+ was due to P2Y receptors. The Ca2+ response was unaltered in the absence of extracellular Ca2+ and strongly reduced by thapsigargin, a blocker of internal Ca2+ stores. The P2Y1 antagonist MRS2179 strongly reduced the ATP- or ADP-induced increase in Ca2+, suggesting the involvement of a P2Y1 receptor. In addition, suramin and PPADS, non-selective antagonists for P2 receptors, inhibited most of the Ca2+ response. The agonistic activity of UTP and the lack of response to UDP implied the additional presence of a P2Y2 and/or a P2Y4 receptors and the absence of a functional P2Y6 receptor. RT-PCR experiments demonstrated that neurospheres expressed P2Y1 and P2Y2 receptors but not P2Y4 receptor. That the majority of the Ca2+ response to ATP was mediated via P2Y1 receptors was also confirmed by analysis of P2Y1 knockout mice and by application of the P2Y1 receptor-specific antagonist MRS2179. In addition, agonists of P2Y1 and P2Y2 receptors and low concentrations of adenosine augmented cell proliferation inspite of the presence of mitogenic growth factors. Neurosphere cell proliferation was attenuated after application of MRS2179 and in neurospheres from P2Y1 receptor knockout mice. These results infer a nucleotide receptor-mediated synergism that augments growth factor-mediated cell proliferation. Taken together these results suggest that P2Y-mediated nucleotidergic signalling is involved in neurosphere function and possibly also in adult neurogenesis in situ.
Active neurogenesis continuously takes place in the dentate gyrus of the adult mammalian brain. The dentate gyrus of the adult rodent hippocampus contains an astrocytelike cell population that is regarded as residual radial glia. These cells reside with their cell bodies in the subgranular layer (SGL). Radial processes traverse the granule cell layer (GCL) and form bushy ramifications in the inner molecular layer (IML). The residual radial glial cells apparently represent neuronal progenitor cells that can give rise to functionally integrated granule cells. To date the cellular and molecular events driving a subpopulation of these cells into neurogenesis as well as the cellular transition states are poorly understood. The present study shows, that in the mouse dentate gyrus, this cell type selectively expresses surfacelocated ATPhydrolyzing activity and is immunopositive for nucleoside triphosphate diphosphohydrolase 2 (NTPDase2). NTPDase2 is an ectoenzyme and hydrolyzes extracellular nucleoside triphosphates such as ATP or UTP to their respective nucleoside diphosphates. The enzyme becomes expressed in the hippocampus during late embryogenesis from E17 onwards, and is thus not involved in early brain development. Its embryonicpattern of expression mirrors dentate migration of neuroblasts and the formation of the primary and finally the tertiary dentate matrix. NTPDase2 is also expressed by a transient population of cortical radial glia from late embryonic development until postnatal day 5. NTPDase2 can be employed as a novel markerfor defining cellular transition states along the neurogenic pathway. It is associated with subpopulations of GFAP and nestinpositive cells. These intermediate filaments are typically expressed by the progenitor cells of the dentate gyrus. In addition there is a considerable overlap with doublecortinand PSANCAM positive cells. The expression of the microtubuleassociated protein doublecortin and of PSANCAM which are expressed by migrating neuroblasts is indicative of a transition of progenitors to a neural phenotype or an immature form of granule cell. NTPDase2 is no longer associated with young neurons and with maturegranule cells, as indicated by the lack of doubleimmunostaining for III tubulin and NeuN, respectively. Furthermore, β S100positive astrocytes do not express NTPDase2 validating that NTPDase2 is also not associated with later stages of gliogenesis. Experiments with the Sphase marker bromodeoxyuridine (BrdU) demonstrate that NTPDase2positive cell proliferate. Postmitotic BrdU-labeled cells preferentially acquire an NTPDase2positive phenotype. Many of these cells were also positive for GFAP. The contribution of BrdUlabeled cells positive for NTPDase2 increased with time from 2 h to 72 h, validating a strong association of NTPDase2 with proliferating cells of the dentate gyrus. The colocalization studies with various markers and the results of the experiments suggestthat NTPDase2 is associated with cell types of varying maturation states but not with mature neurons or astrocytes. Studies on the formation of neurospheres from the dentate gyrus validate previous data suggesting that the hippocampal progenitors have little capacity for self renewal in vitro. In situ hybridization results indicate the presence of one of the metabotropic purinergic receptor subtypes (the P2Y1 receptor) within the adult neurogenic regions, the dentate gyrus and the lateral walls of the lateral ventricles. A patchclamp analysis demonstrates the presence of functional ionotropic nucleotide receptor (P2X receptors) in progenitor cells expressing nestin promotordriven GFP. They suggest that the signaling pathway via extracellular nucleotides and nucleotide receptors may play a role in the control of adult hippocampal neurogenesis.
The physiology of our most complex organ, the brain, is still not comprehensively understood. The brain basically serves the processing, storing and binding of external and internal information, and thereby generates amazing phenomena like the understanding of oneself as an individual entitiy. How exactly information is encoded and represented, how individual neurons or networks of neurons actually interact, is a gigantic puzzle, whose pieces were collected since many decades. Subject of scientific discussions are the basic spatiotemporal structures of neuronal representations. Suggestions and observations reach hereby from simple rate coding of individual neurons to synchronous activity of larger ensembles. To approach answers to these questions, our working group has used a combination of different recording techniques that allowed for the comparison of neuronal interactions on different spatial scales. We focused on prefrontal neuronal interactions during visual short-term memory. Herefore two rhesus monkeys had been trained to perform a visual short-term memory task. We measured and recorded their neuronal activity by means of a microelectrode matrix that could be inserted into the cortex via a closable chamber, which had been previously implanted above prefrontal cortex. The acquired signal was separated into two components: a high-frequency component, that represents the spiking output activity of few neurons in the vicinity of each electrode tip (multi-unit activity), and a low-frequency component, that results from dendritic input activity of larger neuronal assemblies (local field potential). From one of the experimental animals we also recorded mass signals of even larger neuronal populations by means of small silverball electrodes, that had been implated into the skull above prefrontal cortex (skull EEG) in the context of a pilot project. In the first subproject, we analyzed the selectivity of output signals with respect to the memorized stimulus and task performance. We compared selectivities of local recording sites (multi-unit activity) with the selectivities of patterns created by the combined activity of all recording sites, thus representing the activity of large and distributed ensembles. Local neuronal activity correlated with the course of the visual short-term memory task, but was not highly discriminative with respect to different visual stimuli. We could show that the population activity was significantly more specific. Concerning task performance, we obtained the same result, albeit less pronounced. Further analyses revealed that the patterns of distributed ensemble activity were only partly based on realtime coordination of neuronal activity, and in addition, did not remain stable across the time course of the short-term memory task. In the second subproject, we focused on the oscillatory behavior of the local field potential. After a time-frequency analysis, we studied different frequency bands concerning stimulus selectivity and task performance of the monkey. We hereby found significant modulations of oscillations in the beta- and gamma-frequency range, that correlated with different periods of the task. Especially for oscillations in beta- and low-gamma-range, we observed phase-locking of oscillations between different recording sites, which could play an important role as internal clock to coordinate spatially separate activity. Local high-gamma oscillations themselves seemed to be important for the maintenance of information. These results could be partly confirmed by mass signals of EEG. In sum, our results support the hypothesis that information is represented in the brain by means of concerted activity of spatially distributed neuronal ensembles. This activity again appears to be coordinated by oscillatory activity in beta- and low-gamma-frequency ranges. A deeper understanding of central nervous information processing could contribute to better treatment of diseases like Parkinson’s, Alzheimer’s as well as epilepsy, and neuropsychiatric disorders like schizophrenia.
In this thesis, the structure of the C-terminal domain of presenilin-1, the catalytic component of the y-secretase complex, is investigated by NMR spectroscopy. The ysecretase complex has a definitive role in the pathogenic development of Alzheimer's disease, in that it mediates the cleavage of aprecursor to create the amyloid ß peptide. Aggregates of amyloid ß which form amyloid plaques are the most overt clinieal feature observed in the post-mortem brains of Alzheimer's patient. In addition, many of the mutations found in the aggressive early onset familial Alzheimer's disease have been linked to presenilin-1, highlighting its importance in disease progression and deeming it an important target for investigation. One of the greatest challenges for the structural investigation of the y-secretase components is their low expression yields in cell-based systems. We therefore applied continuous-exchange cell-free expression to obtain sufficient amounts of protein for our structural studies. An added benefit of the cell-free expression system is the freedom to incorporate any desired combination of stable-isotope labels directly into sampies. We were therefore able to develop a labeling scheme which targets the amino acid composition of transmembrane a-helices, allowing us to simplify an assignment procedure whieh tends to be cumbersome and diffieult for most a-helical transmembrane proteins. The y-secretase complex is a member of the intramembrane cleaving proteases which, as their name implies, cleave their transmembrane substrates within the bilayer. Single particle analysis of the y-secretase (1) as weil as crystal structures of rhomboid (2) and S2P (3) have revealed the presence of hydrophilie po res within the membrane where catalysis occurs. In light of evidence that certain elements of CTF reside in close proximity or even contribute to the formation of the hydrophilic pore, we chose to study the structure of CTF in mieelles, whieh may be better suited to accommodate CTF in isolation as compared with solid membranes in the absence of the other y-secretase components. The structure of CTF was solved to 1.7 A (backbone r.m.s.d) and revealed the presence of unusual features, including a partially membrane-spanning helix which situates the catalytic asparte at its N-terminus in what would be the center of the membrane where catalysis is proposed to occur, as weil as a severely kinked helix which is partially embedded beneath the surface of the membrane (P6). Interestingly, similar features have been observed in the crystal structure of the GlpG rhomboid. In addition, a soluble helix was found in the long N-terminal loop of CTF which until now has been described as unstructured. The first part of the thesis is designed to provide an introduction to Alzheimer's disease, the role of y-secretase and its presenilin-l catalytic component in disease progression, as weil as cell-free expression and liquid-state NMR techniques involved in the structural investigation of membrane proteins. In chapter 2, the reader is familiarized with the history, the clinical manifestation, and biochemical features of Alzheimer's disease. The chapter goes further to describe the role of the y-secretase complex and its individual components in disease progression and substrate processing. Chapter 3 focuses more specifically on presenilin-l in the context of the newly emerging class of intramembrane proteases. In chapter 4, attention is shifted to the cell-free expression system with special focus on the expression of membrane proteins, and chapter 5 explores the various liquid-state NMR techniques that were required for the characterization of CTF. The second part of the thesis is cumulative and contains original research, method, and review articles that were produced during the course of study. Chapter 6 explores the various techniques and innovations used to study membrane proteins using continuous exchange cell-free expression coupled with NMR spectroscopy. In chapter 7, a new technique, transmembrane segment targeted labeling, is described as a tool that facilitates the backbone assignment of transmembrane proteins which display severe overlap in NMR spectra. Chapter 8 presents the novel NMR structure of the C-terminal fragment of presenilin-l solved in SOS micelles.
The research presented in this thesis characterizes U2AF homology motifs (UHM) and their interactions with UHM ligand motifs (ULM) in the context of splicing regulation. UHM domains are a subgroup of RNA recognition motifs (RRM) originally discovered in the proteins U2AF65 and U2AF35. Whereas canonical RRMs are usually involved in binding of RNA, UHM domains bind tryptophan containing linear protein motifs (ULM) instead. In the first article, we analyze the complex network of interactions between splicing factors and RNA that initiate the assembly of the spliceosome at the 3´ splice site of an intron. The protein U2AF65 binds a pyrimidine-rich element in introns and recruits U2snRNP by binding its protein component SF3b155. My contribution was to define the binding site of the protein U2AF65 to the intrinsically unstructured N-terminus of the scaffolding protein SF3b155. I could show that the UHM domain of U2AF65 recognizes a ULM in SF3b155, and that this binding site is not overlapping with the binding sites of other splicing factors, like p14, to SF3b155. As the U2AF65-UHM:SF3b155-ULM interaction is mutually exclusive with an interaction between U2AF65-UHM and a ULM in the splicing factor SF1, which was reported to initially recognize the branch point sequence, my results provide the molecular details on how SF3b155 replaces SF1 during spliceosomal reorganizations. In the second article, we show that overexpression of the UHM domain of the splicing factor SPF45 induces exon 6 skipping in the pre-mRNA of Fas (CD95/APO-1). I provide evidence for in vitro binding of SPF45-UHM to ULM sequences in the splicing factors U2AF65, SF1, and SF3b155. I crystallized free and SF3b155-bound SPF45 UHM and solved both structures by X-ray crystallography. The analysis of the complex interface and sequence differences in the ULMs allowed me to design mutations of SPF45-UHM, which selectively inhibit binding to distinct ULMs. After assessing the ULM binding properties in vitro, we could show that the activity of SPF45-UHM in influencing the splicing pattern of Fas relies on interactions with SF3b155 and/or SF1, but that an interaction with U2AF65 is dispensable. A mechanism for the activity of SPF45-UHM could thus be engaging in ULM interactions and thus interfering with the network of interactions that initiate the assembly of the spliceosome at the 3´splice site, as described above. In the third article, we describe an unusual flexible homodimerization mode of the UHM in the splicing factor Puf60, which enables simultaneous interactions with ULM sequences on other splicing factors. I could show that the NMR relaxation properties of Puf60-UHM are inconsistent with a model of a rigid dimer, but rather indicate a dimerization via a flexible linker. I identified a flexible loop in the peptide backbone of Puf60-UHM, and showed that mutiation of acidic residues in this loop impairs the dimerization. To analyze the dimerization interface in further detail, I solved the structure of Puf60-UHM by X-ray crystallography. The acidic residues in the flexible loop of one UHM dimer subunit mediate the dimerization by contacting basic residues on the β-sheet surface of the other dimer subunit. Differences in the four dimer interfaces observed for the eight molecules in the asymmetric unit of the crystal support the model of an undescribed, flexible mode of dimerization, and thus complement the NMR relaxation data. Furthermore, I could show that the Puf60-UHM dimer and U2AF65-UHM contact different ULM sequences on the SF3b155 N-terminus in vitro, thus providing a possible explanation for the mutual cooperative activation of Puf60 and U2AF65 in splicing assays described in the literature. The fourth article is a review about recent research on the recognition of DNA double strand breaks (DSB) by covalent histone modifications. The p53 binding protein 1 (53BP1) is a DSB sensor and a checkpoint protein for mitosis. Recent crystallographic evidence indicates that 53BP1 recognizes DSB sites by binding histone H4 dimetylated at lysine 20 (H4-K20). We provide a comprehensive overview of the atomic resolution structures that revealed how proteins can specifically recognize histone tail modifications, especially methylated lysines, to read the information stored in what is called the histone code.
(1) Die genomweite Expressionsanalyse von salzadaptierten Zellen von M. mazei Gö1 identifizierte eine Reihe von salzregulierten Genen. Neben den beiden Operone ota und abl, die für die Akkumulierung von Glycin-Betain und Ne-Azetyl-b-Lysin verantwortlich sind, konnte ein ABC-Transporter (MM0953), der in seiner Genumgebung weitere Transporter sowie Proteine mit konservierten S-Layer-Domänen aufweist, als salzreguliert erkannt werden. Dies deutet auf ein S-Layer-Exportsystem hin, das eine Rolle in salzadaptierten Zellen spielen könnte. (2) Eine genomweite Expressionsanalyse von Zellen von M. mazei Gö1 zu unterschiedlichen Zeitpunkten nach einem hyperosmotischen Schock auf 400 mM NaCl ermöglichte Einblicke in den Verlauf der Genexpression. Die Erhöhung der externen Osmolarität resultierte in der erhöhten Expression von Genen, die für die Aufnahme und Biosynthese von kompatiblen Soluten verantwortlich sind sowie von Genen deren Produkte regulatorische Funktion haben könnten. (3) Genomweite Expressionsanalysen von Zellen von M. mazei Gö1 nach einem hypoosmotischen Schock zeigten erhöhte Expression von Genen, die an der Regulation und an der generellen Stressantwort beteiligt sind. Gene, deren Produkte im Stoffwechsel wichtig sind – besonders Gene, die für Methylamin-Corrinoid-Methyltransferasen kodieren – erscheinen stark reprimiert. (4) Die Bestimmung der intrazellulären Ionenkonzentrationen zeigte ein unspezifisches Einströmen von den Ionen, die den osmotischen Schock auslösen sofort nach dem Schock, sowie den Ausstrom derselben Ionen im Verlauf von 5 Minuten. Die Ionenkonzentrationen der Ionen, die den Schock auslösten, blieben intrazellulär erhöht. Das Ein- und Ausströmen der Ionen nach einem hyperosmotischen Stress ist nicht energieabhängig. (5) M. mazei akkumulierte nach einem hyperosmotischen Schock kein K+, zeigte aber eine erhöhte intrazelluläre Konzentration dieses Ions, wenn die Zellen in Medium mit erhöhter Osmolarität angezogen wurden. (6) Durch hyperosmotische Schocks mit verschiedenen Salzen und Zuckern konnte gezeigt werden, dass die kurzzeitige Akkumulation von Ionen keine gerichtete Antwort auf den osmotischen Stress ist. (7) Es konnte weiters gezeigt werden, dass Zellen von M. mazei Gö1, die mit dem kompatiblen Solut Betain inkubiert wurden, nach einem hyperosmotischen Schock K+ akkumulieren. Dies bedeutet möglicherweise eine K+-abhängige Regulation des Glycin-Betain-Transporters. (8) Die Funktion der drei im Genom kodierten Na+/H+-Antiporter konnte auf transkriptioneller Ebene nicht geklärt werden. Trotzdem zeigt ein Hydrophobizitätsplot des Proteins eine mögliche Beteiligung von Nha1 (MM0294) an der Osmoregulation durch eine hydrophile C-terminale Domäne. (9) Nach einem hyperosmotischen Schock von 38,5 auf 400 mM NaCl erhöhte sich die intrazelluläre Konzentration an Glutamat, das in M. mazei als kompatibles Solut fungiert, bereits nach drei Stunden. Zellen, die bereits an die erhöhte Salzkonzentration adaptiert waren, enthielten 1,4 μmol Glutamat/mg Protein. (10) Die Glutamin-Synthetase zeigte eine erhöhte Transkription nach einem hyperosmotischen Schock. Das Protein wird aber nicht salzabhängig produziert und zeigt keine Enzymaktivität. Die Biosynthese des Solutes über eine Glutamat-Dehydrogenase ist die wahrscheinliche Alternative. (11) Aufgrund der generierten Expressionsprofile und der physiologischen Daten konnte ein Modell der Osmoadaptation in Methanosarcina mazei Gö1 erstellt werden.
Paleoecology is the study of organismal interactions with the environment in the geological past. Organisms are influenced in their distribution and abundance by abiotic factors such as temperature and precipitation. A change in these factors, for example by major climatic shifts, would then affect the communities of organisms. Studying this hypothesized causal link between climatic and faunal change is especially interesting for the Plio-Pleistocene of East Africa due to the fact that our own ancestors also inhabited these regions. Both the Turkana basin in Kenya and the Lake Albert region in Uganda offer unique opportunities to investigate these paleoecological issues. Their late Miocene through Pleistocene deposits provide a very good record of climatic, vegetation and faunal change in East Africa (Pickford et al. 1993, Leakey et al. 1995, 1998, McDougall & Feibel 2003, Wynn 2004). This study focuses on the mammal family Bovidae as they are good indicator of vegetation and environment (e.g. Vrba 1980, 1995, Shipman & Harris 1988, Bobe & Eck 2001, Bobe & Behrensmeyer 2004, Bobe et al. 2007). Bovidae are quite species-rich and inhabit a wide range of habitats from tropical rain forests to deserts which predicates their array of morphological adaptations (ecovariables) to these environments. Diet is the ecovariable that is most to climate and thus habitat change. Therefore, the fossil Bovidae are especially suitable for reconstructing past environments. The objective of this thesis is to test the hypothesis that, from the late Miocene through the Holocene, Africa has experienced an overall increase in aridity and concomitant pulses of habitat change. The hypothesis predicts that increasing aridity causes a likewise growth in the abundance of taxa adapted to open arid environments. In particular, an increase in bovid grazers should be observed in combination with a decrease of bovid browsers. To test this hypothesis, I examine the fossil bovid communities from each stratigraphic member of Lake Turkana (Lothagam, Kanapoi, West Turkana and Koobi Fora) and Lake Albert (Nkondo-Kaiso region) and through a taxonomic and a functional perspective reconstruct the paleoenvironments and -climates from approximately 8 to 0.6 Ma. This study is the first to use taxonomic and ecomorphological data together to reconstruct the paleoenvironments of the Turkana basin and the Nkondo-Kaiso region of Lake Albert. In a first analysis, mesowear, as introduced by Fortelius & Solounias (2000), is used to gather information about the diet of bovids. As a result of my preliminary investigations on upper vs. lower molars of recent species, the sample of fossil bovid specimens from the Turkana basin and Lake Albert were found to be unsuitable to reveal a meaningful diet reconstruction. Therefore, the bovids are assigned to diet categories based on literature. For each member of the time period from 8.0 to 0.6 Ma, I provide a detailed characterization of the bovid fauna in terms of α- and β- diversity both on tribe and diet level based on presence-absence as well as for the Turkana basin on abundance data. Statistical comparisons between the fossil bovid communities and those in modern protected areas with known vegetation and climatic conditions have yielded modern analogues for each stratigraphic member. Following that I provide paleoclimatic conditions such as assumed mean annual temperature for each member. Based on abundance of diet categories in the bovid communities, the paleoclimate of the Turkana basin was in general cooler and considerably more humid during the late Miocene to the Pleistocene than today. The mean annual temperature at Lothagam is assumed as 22.2 °C, the annual precipitation as 685 mm for 8.0 – 6.54 Ma and 4.9 – 3.4 Ma. The intervening time period is characterized by a slightly lower mean annual temperature and precipitation (20.3 °C, 583 mm). From 4.17 to 4.07 Ma Kanapoi faced 21.3 °C and 592 mm rainfall. In the eastern part of the basin the climate was warmer and more humid (3.4 – 2.68 Ma: 26.2, 961 mm; 2.68 – 1.3 Ma: 27.1 °C, 935 mm) from 3.4 to 1.3 Ma than in the preceeding eras. In the western part, the climate became warmer and more humid ~500,000 years later and was more variable than that in the eastern basin. From 2.94 to 2.52 Ma the mean annual temperature was 26.2 °C and the annual precipitation 961 mm. Between 2.34 and 1.6 Ma the climate again cooled and became drier as before 2.94 Ma. A second shift to higher temperature and precipitation occurred after 1.6 Ma (27.1 °C, 935 mm) lasted until 1.34 Ma. The results of the bovid community analyses do not support the hypothesis of increasing aridity in Eastern Africa during the late Mio- to Pleistocene. Instead, the results show that the bovid communities differed much over time and on a relatively small spatial scale. Regional paleovegetation and paleoclimate exhibit fluctuations through the studied time period at western Turkana and differences between the western and eastern part of the Turkana basin. This is indicative of a patchy habitat distribution both on temporal and spatial levels. Increased climate variability predicts an increase in landscape complexity as proposed by the ‘variability selection hypothesis’ (Potts 1998a+b). Therefore, this thesis research supports the hypothesis of increased landscape complexity on the spatial level. This study has important implications for future research. First, an analysis based on ecovariable characteristics such as diet may be preferred to a taxonomic analysis. Second, abundance data should be used for an ecovariable analysis because the results then provide more precise information on the paleovegetation and –climate than just the presence of these adaptations in the faunal community. Lastly, as this study is based on one mammal family, further studies on other mammal groups should be conducted to increase the database of exploited resource by the entire faunal community. Most significantly this study provides a basis for new interpretations of faunal community distributions. It also raises the question whether small scale spatial community variability is also to be expected at other fossil sites. If so then this methodology has important implications for reconstructions of paleovegetation and paleoclimate.

Ginkgo biloba (Gb), eine der am besten untersuchten pharmazeutisch-medizinisch genutzten Pflanzen, wird heute in Form von Spezialextrakten im Sinne einer evidenzbasierten Phytopharmakatherapie eingesetzt. Grundlage hierfür sind die genaue Spezifikation der Zusammensetzung des Spezialextraktes in Bezug auf die wirksamkeitsbestimmenden Inhaltsstoffe, balstbare klinische Daten, das Erforschen des molekularen Wirkmechanismus‘ des Gesamtextraktes aber auch der Einzelbestandteile und die Pharmakokinetik im Targetgewebe. Heute werden im Sinne einer evidenzbasierten Phytopharmakatherapie lediglich Extrakte verwendet, die der Monographie der Komission E entsprechen (22 - 27% Flavonoide, 5 - 7% Terpenlaktone und weniger als 5 ppm Ginkgolsäuren). Der am besten klinisch und pharmakologisch untersuchte Gb-Spezialextrakt ist EGb 761® (Tebonin®), der im zentralen Fokus der vorliegenden Arbeit steht. Die im Jahr 2008 vom IQWiG veröffentlichte Metaanalyse zur Klinik von EGb 761® hat in äußerst detaillierter Form belastbare Daten zur Wirksamkeit dieses Extraktes beschrieben. Es kann festgehalten werden, dass ein Einsatz dieses Spezialextraktes im Rahmen der Therapie einer beginnenden Demenz zu befürworten ist. Basis des klinischen Einsatzes des EGb 761® sind in vitro und in vivo pharmakologische Untersuchungen. Es werden unterschiedliche Gesamtkonzepte zur Wirkung von EGb 761® bzw. Einzeleffekte der Inhaltsstoffe im ZNS diskutiert. Konsensfähig sind heute sicher die Mitochondrien-stabilisierende Wirkung der Terpenlaktone und ein antioxidativer Effekt der Flavonoide. Bb zeigt zusätzlich deutlich protektive Effekte in Bezug auf durch zerebrovaskuläre Ereignisse geschädigte Hirnareale. Darüber hinaus ist die Wirkung der Flavonoide auf die monoaminerge Neurotransmission aktueller Gegenstand der Forschung. Basis jeglicher pharmakologischen Betrachtung ist das pharmakokinetische Verhalten der wirksamen Inhaltsstoffe im Target-Gewebe. Nachdem die ZNS-Bioverfügbarkeit der Flavonoide nachgewiesen wurde, hat die vorliegende Arbeit das zentrale Ziel, die pharmakokinetische Charakteristik der Terpenlaktone aus Gb im ZNS zu untersuchen. Zur quantitativen Analyse der Terpenlaktone (GKA, GKB, GKC und Bb) in biologischen Matrices (Hirn-Homogenat, Plasma) und Hirn-Dialysat-Pufferlösung (aCSF-Puffer) wurde eine LC-MS-Analytik-Methode entwickelt und validiert. Unter Verwendung einer 250x4 mm, Multo High 100 RP18, 5 μm (CS-Chromatographie Service GmbH)-Säule und einer isokratischen Auftrennung mittels einer mobilen Phase bestehend aus 60% 0,1%-iger Ameisensäure und 40%-igem Methanol konnten alle vier genannten Terpenlaktone simultan innerhalb von 20 Minuten analysiert werden. Die beschriebene LC-MS(TOF)-Methode verfügt über eine ausreichende Sensitivität, um die Analyten im nanomolaren Bereich zu quantifizieren (z.B. LOQ Bb in aCSF-Puffer: 0,25 pg/μl; LOQ Bb in Hirnhomogenat: 1 ng/ml). Die Aufarbeitung der Plasma- bzw. Hirn-Homogenat-Proben erfolgte durch eine flüssig-flüssig-Extraktion mit Hilfe von Extrelut®-Säulen; die Hirn-Dialysat-Proben bedurften keiner Probenaufarbeitung. Mit Hilfe der beschriebenen Analytik-Methode war es möglich, GKA, GKB, GKC und Bb in Plasma und Hirnhomogenat von Ratten nach oraler Gabe von 600 mg/kg Körpergewicht EGb 761® bzw. einer vergleichbaren Menge der Reinsubstanzen zu bestimmen. Im Rahmen dieses Projektes wurde ein direkter Vergleich der erhalten Plasma-Konzentrationen nach Extrakt- bzw. Reinsubstanzgabe gezogen, wobei der Extrakt die höhere AUC (für GKA u. Bb) und daher bessere Bioverfügbarkeit aufwies. Es konnten in Plasma und Gehirngewebe sowohl GKA als auch GKB und Bb in nativer nicht metabolisierter Form nachgewiesen werden. GKC konnte weder in Plasma noch in Hirngewebe bestimmt werden, was die in der Literatur diskutierte These einer schnellen Metabolisierung (Methylierung) stärkt. Die Terpenlaktone sind im Plasma sehr schnell angeflutet und zeigten ein ebenfalls zügiges Abfallen, so dass 24 Stunden nach oraler Applikation keine Konzentrationen mehr zu detektieren waren. Bei der Untersuchung der Hirn-Gewebspiegel von GKA, GKB und Bb zeigten sich keine Unterschiede nach Gabe von Extrakt bzw. Reinsubstanz. Die Substanzen fluteten im Vergleich zum Plasma etwas verzögert an, fielen aber auch bis 24 Stunden nach Applikation wieder unter die Nachweisgrenze. Die Konzentrations-Zeit-Kurven ähnelten in ihrer Form stark denen aus Plasma, waren jedoch zeitlich nach rechts verschoben, so dass ausgeschlossen werden kann, dass es sich im Hirngewebe um Artefakt aus Restblut handelt. Wesentliches Resultat dieser Untersuchungen war, dass erstmalig nach oraler Gabe von EGb 761® gezeigt wurde, dass deutliche Gewebespiegel im Gehirn von Ratten zu erzielen sind und damit diese Substanzen im Target-Gewebe die postulierten pharmakologischen Wirkungen ausüben können. Aufbauend auf diesen Ergebnissen wurden mit Hilfe der Mikrodialyse-Technik und der bereits beschriebenen LC-MS-Analytik-Methode weitere pharmakokinetische Untersuchungen am Maus-Modell durchgeführt. Es konnte zunächst rein technisch im Rahmen von Wiederfindungsuntersuchungen gezeigt werden, dass die im Dialysat bestimmte Menge Bb ca. 6% der tatsächlich im Extrazellularraum des Maus-Hirns vorliegenden Bb-Konzentration entspricht. Weiterhin zeigten diese Versuche, dass Bb kaum an Plasma-Proteine bindet, da keine signifikanten Unterschiede bei der Dialyse von Bb aus Puffer, Blut oder Plasma zu sehen waren. In einem ersten Tierversuch an gesunden Mäusen konnten die pharmakokinetischen Charakteristika von Bb, die in der Fütterungsstudie an Ratten bestimmt wurden, reproduziert werden, obwohl es sich um einen völlig divergenten Versuchsaufbau, unterschiedliche Tierspezies und nicht um die gleichen Applikationsformen handelt. Diese Tatsache unterstreicht die Aussage beider Studien. Als zusätzliche Aussage ergibt sich aus dem Versuchsaufbau, dass Bb frei und biologisch aktiv im Extrazellularraum vorliegt und nicht z.B. in Membranen gebunden ist. Die Möglichkeit mittels Mikrodialyse und LC-MS-Technik Bb im Extrazellularraum definierter Hirnregionen nachzuweisen, erlaubte eine pharmakokinetische Charakterisierung von Bb in vom Schlaganfall geschädigten Hirngewebe. Es zeigte sich, dass bei Gabe von 10 mg/Kg Bb eine Stunde vor dem Schlaganfall die Bb-Konzentrationen zwar deutlich abfallen, aber dann relativ konstant bleiben, was durch einen fehlenden Abtransport durch die unterbrochene Blutversorgung zu erklären ist.
Phykologie - Blütezeiten und Ruhepausen einer Wissenschaft im Nebenamt (Fallbeispiel Grünalgen)

Phykologie (Algenforschung) ist der Öffentlichkeit dem Namen nach und als wissenschaftliche Disziplin kaum geläufig. Innerhalb der Botanik (Biologie) besteht die Algenkunde gleichsam "implizit" als eine Vielfalt von Entwicklungssträngen, die selten gebündelt sind, vielmehr leicht immer wieder abreißen können, denen sich aber immer wieder auch neue Stränge hinzugesellen. Die Phykologie erforscht, im Gegensatz etwa zum Studium der Blütenpflanzen, nur wenige aus dem Alltagsleben vertraute Gewächse. Ihre Untersuchungsobjekte wurden (und werden immer noch) erst nach und nach entdeckt, weshalb sich die Vorstellung, was eine Alge ist, ständig wandelt. Viele sind ohne Mikroskop gar nicht wahrzunehmen. Forschungsgegenstand und Forschungsaufgabe bilden durch ihre fortwährende Umgestaltung die Leitgedanken der Botanik und Biologie überhaupt ab, wobei die Beziehung reziprok ist. Eine lebhafte Fluktuation von ausübenden Personen und Paradigmata ist ebenso charakteristisch für die Phykologie wie eine ausgeprägte Inter- bzw. Transdisziplinarität und ein hohes aktives internes Innovationspotential. Am Beispiel der traditionellen, heute so nicht mehr bestehenden, »Chlorophyta« wird untersucht, wie diese Sippe zunächst für makroskopisch sichtbare Meeresalgen etabliert wurde. Dann wurden ihr schrittweise verschiedene binnenländische Formenkreise von Algen zugeordnet, bis hin zu den grünen Phytoflagellaten. In der jüngsten Entwicklungsphase haben sich die "grünen Algen" als heterogener Komplex mehrerer voneinander unabhängiger Entwicklungslinien erwiesen, die bis auf eine über das Niveau der Algen nicht hinausgekommen sind, wohl aber zum Landleben fähige Vertreter haben. Als Wurzelbereich der echten Landpflanzen erwies sich überraschend ein kleiner Verwandtschaftsbereich der Algenklasse der Charophyceae.
This study comprises a survey on ecology, morphology and taxonomy of parasitic fungi infecting Pteridophytes and Orchidaceae found by the author on several field trips to Western Panama as part of the project plant parasitic micro-fungi of Western Panama (ppMP). In Panama, approximately 9500 species of vascular plants are found. Of these, Orchidaceae are with ca. 1150 (ca. 12%) species by far the most speciose family. The Pteridophytes in Panama comprise ca. 940 species in 31 families. Most fungal pathogens on Orchidaceae in tropical regions were described from plants in culture or from material intercepted at borders by plant quarantine services and not from their natural habitats. Therefore, little is known about distribution and ecology of these pathogens in their natural range. The author determined and classified several hundred Orchidaceae-species and Pteridophytes at the sites selected in the context of the project. This work facilitated the identification of many host plants (at least to genus-level) even in sterile condition in the field. About 65 species of Pucciniales are known to infest Orchidaceae and ca. 38% of them are described from tropical America. All available types of Pucciniales on Orchidaceae in tropical America were studied and compared with 91 specimens of rust fungi on orchids collected by the author in Panama. Several hundred additional specimens housed in the BPI, almost all intercepted from plant quarantine services, were used for comparison. As result of this work, it is suggested to combine Uromyces stenorrhynchi Henn. to Sphenospora and, as this is the oldest epithet, to synonymize S. kevorkianii Linder, S. mera Cumm. and S. saphena Cumm. with it. Further, it could be demonstrated that Uredo aurantiaca Montemartini, U. cyrtopodii Syd. & P. Syd., U. epidendri Henn., U. guacae Mayor, U. gynandrearum Corda, U. lynchii (Berk.) Plowr., U. neopustulata Cumm. (≡U. pustulata Henn.), U. nigropuncta Henn., U. oncidii Henn., U. ornithidii F. Kern., Cif. & Thurst., and presumably U. scabies Cke., are anamorphs of this variable species. U. gynandrearum is the oldest anamorph-name for all these taxa. Therefore, it can be established that this rust infects more than 80 species of Orchidaceae in three subfamilies. In total, the anamorph of this species was collected by the author on 17 different species of Orchidaceae in Panama which, apart from one species, are all new hosts to science. The molecular data obtained by the author confirm this view, although more data, especially from material from the whole range of distribution of U. gynandrearum, are necessary. Puccinia spiranthicola Cumm. was found to be a synonym of P. cinnamomea Diet. & Holw. and was found by the author on three different Orchidaceae in two subfamilies. Uredo pleurothallidis Keissl. is now considered a synonym of U. wittmackiana Henn. and the latter as the anamorph of Puccinia oncidii Cumm. In the anamorph genus Uredo, a new species was found infecting at least five different species of Sobralia and Elleanthus (Sobraliinae) at different localities. Molecular data indicate it to be related to the currently polyphyletic Phakopsoraceae. For the rusts with suprastomatal sori on Orchidaceae, now separated from Hemileia and placed in the genus Desmosorus (nom. inval.), the current concept with only one taxon is rejected and the establishment of three subspecies is suggested. The complicated taxonomy is discussed and makes it necessary to validate the genus-name and make a new combination. Another Hemileia-anamorph species was found by the author and is considered to be new to science. This is the first species of this alliance in America on Orchidaceae. Molecular data obtained by the author confirm the separation of Desmosorus from Hemileia and the position of the new species. For rusts on Pteridophytes, a new species of Milesia, (teleomorph: Milesina) and a new anamorphic species of Uredinopsis was found, both on hosts hitherto not known. In Calidion, the presumable anamorph-genus of Uncol, the species C. cf. cenicafeae Salazar & Buriticá was found on several new hosts. Further, the teleomorph was found. Morphologically, this teleomorph did not agree with the description of Uncol by the author of the genus, although the anamorph characteristics left no doubt that it is Calidion. Apparently, the description of Uncol is inadequate, but cannot be improved, as the type is unavailable. Molecular data obtained by the author show this species to be closest to Desmosorus. For Uredo superficialis Speg., the anamorph of Desmella, nine new hosts in eight different fern families were found by the author and the collaborators of the ppMP-project. Ecological data indicate that this species includes different host specific races, which, however could not be distinguished morphologically. For all these rusts, a thorough discussion of the ecology in their habitats is given. In total, 21 LSU rDNA sequences from 6 different rust species on Orchidaceae and Pteridophytes were obtained and analyzed with the Maximum Parsimony and Minimum Evolution method. Here, the position of several groups could be confirmed, and some anamorphs could be assigned to different teleomorphic relationships. Within the Ascomycota and their anamorphs, several hitherto unknown species and species not known from these hosts or not known from Panama were found and analyzed. On Orchidaceae, the following fungi belonging to the Ascomycota are described, illustrated and discussed: In the Phyllachorales, a hitherto not known Phyllachora sp. was found on Oncidium warszewiczii Rchb. f. and was compared with the other species of this order currently known from Orchidaceae. In the Asterinaceae s. l. Lembosia cf. epidendri Meir. Silva & O. R. Pereia was found on Maxillaria crassifolia (Lindl.) Rchb. f., which is a new host and new host alliance for this fungus hitherto only known from Brazil. The fungus is described and compared with all species of Asterinaceae currently known on Orchidaceae. In the Meliolaceae, Meliola orchidacearum Cif. was found on Camaridium biolleyi (Schltr.) Schltr. and an Epidendrum sp. which are new hosts and new host alliances of this fungus which was hitherto only known from the Caribbean Islands. It is described, illustrated and compared with the type. In the Glomerellaceae, Glomerella cingulata and its anamorph Colletotrichum gloeosporioides were found on several hosts. The species is illustrated, described and compared with data from literature. In the anamorphic Mycosphaerellaceae, Pseudocercospora odontoglossii (Prill. & Delacr.) U. Braun, a species currently only known from culture, was found on the new host Pleurothallis imraei Lindl. It is illustrated, described and compared with data from literature. On ferns, the following other fungi are described, illustrated and discussed: A conspicuous undescribed form of Polycyclus was found by the author on Elaphoglossum ciliatum (C. Presl.) T. Moore (Dryopteridaceae) and Serpocaulon loriceum (L.) A. R. Sm. (Polypodiaceae). A conspectus of Parmulariaceae infecting ferns is given and demonstrated that Polycyclina should be synonymized under Polycyclus. Summing up, it can be assessed, especially for the Pucciniales, that the most speciose plant family in Panama carries remarkable few species of specific parasites, and that many of them seem to be distributed over a wide range of species which often are not closely related. One reason amongst others seems to be that parasites need a minimum density of host plants in a habitat to survive. As orchid species often occur with only few (and often small) individual plants at a given locality, the probability for a specific pathogen to infect a plant gets too low, hence high diversity by low abundance of hosts might be an impediment for specific pathogens. In this case, unspecific parasites, or such which are infecting larger alliances, are in advantage. Other reasons could be specific traits of orchids, like succulence and mycotrophy which might hamper fungal infections.
5-Lipoxygenase (5-LO) catalyzes the two initial steps in the biosynthesis of leukotrienes, a group of inflammatory lipid mediators derived from arachidonic acid. Here, the regulation of 5-LO mRNA expression by alternative splicing and nonsense-mediated mRNA decay (NMD) was investigated. In the present study, the identification of two truncated transcripts and four novel 5-LO splice variants containing premature termination codons (PTC) was reported. The characterization of one of the splice variants, 5-LOΔ3, revealed that it is a target for NMD since knockdown of the NMD factors UPF1, UPF2 and UPF3b in the human monocytic cell line Mono Mac 6 (MM6) altered the expression of 5-LOΔ3 mRNA up to 2-fold in a cell differentiation-dependent manner suggesting that cell differentiation alters the composition or function of the NMD complex. In contrast, the mature 5-LO mRNA transcript was not affected by UPF knockdown. Thus, the data suggest that the coupling of alternative splicing and NMD is involved in the regulation of 5-LO gene expression.
RT-PCR analysis of different cell types revealed the existence of a large number of 5-LO splice variants. The most interesting splice variants were observed in BL41-E95A cells, which give a raise to novel 5-LO protein isoforms. This leads to the hypothesis of a novel regulatory mechanism in which the dimerization of 5-LO with 5-LO isoforms might regulate the 5-LO activity.
The 5-LO protein expression was reduced on translational level in UPF1 knock down cells, suggesting that UPF1 has a positive influence on 5-LO translation. Therefore, a mass spectrometry based proteomics study was started to identify compartment specific protein expression changes upon UPF1 knockdown in differentiated and undifferentiated MM6 cells. The proteomics analysis demonstrated that the knockdown of UPF1 results in numerous protein changes in the microsomal fraction (~ 21%) but not in the soluble fraction (< 1%). Western blot data confirmed the trend of the proteomics analysis. This data suggest that UPF1 is a critical gene expression regulator in a compartment specific way. During differentiation by TGFβ and calcitriol the majority of UPF1 regulated proteins was adjusted to normal level. It appears that that not only the NMD mechanism alters its composition during differentiation. Also the gene expression regulation on translational level by UPF1 seems to be also cell differentiation dependent. An interesting group of UPF1 target genes represent the downregulated proteins. qRT-PCR analysis of randomly chosen genes revealed no effect on mRNA expression upon UPF1 knockdown, suggesting that UPF1 positively influences the translation of these genes. Computational sequence analysis identified a conserved C-rich sequence which might be a hnRNP E2-binding site. hnRNP E2 has been characterized as a translational repressor in myeloid cells. Western blot analysis revealed a differentiation independent up regulation of hnRNP E2 by UPF1 knockdown. Additionally, microRNA-328 (miR-328) has been described as an RNA decoy modulating hnRNP E2 regulation. Due to this, stem loop qRT-PCR showed an up regulation of miR-328 in TGFβ and calcitriol differentiated MM6 cells. Based on this data we suggest a model in which downregulation of UPF1 increases hnRNP E2 expression, leading to translation inhibition. During differentiation, miRNA-328 is upregulated thereby competing with hnRNP E2 leading to an efficient translation
Since combinatorial chemistry and high throughput screening have become a common technique in the drug discovery phase the number of compounds being considered has increased frequently. These structures are often characterized by high molecular weight, high lipophilicity and low solubility in aqueous and physiological media. Due to the generally poor bioavailability, new in vitro techniques were needed for screening of pharmacokinetic properties. An important parameter for these screening methods is the implementation at an early state of drug discovery phase, to find potential lead structures, before investment costs become significant. The established in vitro methods for the prediction of membrane interaction are not reliable especially for poorly soluble compounds. A new method that is fast and easy to use, requires only small amounts of NCE and which can provide more reliable predictions is needed. In this study, a new screening technique based on surface activity profiling for the prediction of oral drug absorption was evaluated with special emphasis on the predictability of biological membrane interaction of poorly soluble drug compounds. It was demonstrated that drug absorption through a bilayer membrane can be modeled by the orientation of compounds at the air/water interface. Thus amphilicity of a drug is generally related to both oral absorption and blood brain barrier penetration. In turn, amphiphilicity is influenced by the lipophilicity, size and charge distribution of a drug. Surface activity profiling was determined by analysis of surface pressure profiles using the Gibbs adsorption isotherm. The surface activity measurements were carried out using a multichannel tensiometer Delta 8, which was developed by Kibron to be utilized in conjugation high throughput screening in early drug discovery processes. For this study two test sets were analyzed, one for the prediction of gastrointestinal wall interaction and the second for the prediction of the penetration behavior at the blood brain barrier. Both test sets consist of drug compounds with a wide range of absorption properties and consist mainly of compounds with poor water solubility. Since the drugs characteristics varied, they were classified according to water solubility and surface activity and a sample preparation method for each group was established. For the prediction of oral drug absorption, three different methods were established to model the interaction of compound and gastrointestinal wall. For drug compounds with solubility above 1mmol/L the traditional shake-flask method enabled the determination of the amphiphilic properties of drug compounds in pure aqueous media. Compounds with solubility below 1mmol/L tend to not to exhibit any increase in surface activity. Thus surface tension measurements of compounds, which exhibited a limited surface activity due to poor aqueous solubility, were conducted from stock solutions prepared with various organic solvents. Mainly polar organic solvents were used. A mixture of DMSO and DMF resulted in the best combination of properties: the intensive solubility enhancing effect of DMF and the lower intrinsic surface activity of DMSO. The polar solvent ruptured the water clusters, so that highly lipophilic structures had a higher affinity to the solvent and higher concentrations could be obtained. For these compounds higher maximum surface pressure were generated than was possible in pure aqueous media. The surface pressure data were correlated with the fraction absorbed values in vivo. However it was found that poor water solubility is not the only limiting step to exhibiting any surface activity. Some compounds were showed no surface activity in either solvent system. Therefore a micelle vehicle method was established using short chain phospholipids to mimic the gastrointestinal wall. It could be concluded from the results, that non surface active drugs can interact with the phospholipids micelle vehicle in a way analogous to their interaction with the membrane bilayer. The relative critical micelle concentration was correlated with the fraction absorbed of this test set. A sample preparation schema based on the three types of drugs was established. This schema enabled us to predict the absorbance of slightly soluble and poorly soluble drugs with acceptable reliability for early compound screening. For the prediction of blood brain barrier penetration using surface activity profiling as analyzing method, a test set with very poorly soluble characteristics was chosen. The sample preparation method was based on a strictly aqueous approach using the ‘shake flask’ method. The surface tension measurements enabled correlation of the amphiphilic properties of the very poorly soluble drug compounds with BBB uptake. From the aqueous surface pressure profiles and the determination of physicochemical parameters, it was found that blood brain barrier is more likely when a drug provides a small cross-sectional area, As, at the interface. The cross-sectional area is the only parameter which is independent from the maximal concentration in aqueous media and it is particularly suitable for lower solubility compounds. In summary, it was shown that amphilicity is related to biological membrane interaction in the human body and that surface activity profiling with appropriate sample preparation can be used as a reliable screening tool for the prediction of oral drug absorption of poorly soluble drugs. Furthermore an in vitro screening method of blood-brain-barrier penetration was established.
Summary:
1) Three types of forest, evergreen seasonal forest, heath forest and Melaleuca swamp forest, were distinguished and studied in the vicinity of Cheko in southwestern Cambodia, where moist tropical climate with a pronounced dry season in three winter months prevails.
2) These three forest types respectively occupied deep latosol derived from sandstone, very sandy soil around the swamp forest, and deep deposit of silica sand with underground hardpan in shallow valleys.
3) Total plant biomass was estimated by the allometric method based on some 140 sample trees (DBH24.5 cm) which were felled in four sample plots (two 50 mX50 m plots in the evergreen seasonal forest, and each one 20 m x 50 m plot in the other two types). Biomass of ground vegetation was estimated separately by similar technique and clipping.
4) The biomass of evergreen seasonal forest was estimated as follows. Stem 215 ton/ha, branch 99 ton/ha, root 61 ton/ha, leaf 7.3 ton/ha, leaf area index 7.4 ha/ha, density of trees over 4.5 cm DBH 1,280/ha, relative basal area of Whole stand 3.19 o/oo.
5) The biomass of heath forest was as follows. Stem 111 ton/ha, branch 35 ton/ha·, root 19 ton/ha, leaf 7.7 ton/ha, leaf area index 7.1 ha/ha, tree density 2,570/ha, relative basal area 2.3 o/oo.
6) The biomass of M elaleuca swamp forest was as follows. Stem 7.4 ton/ha, branch 3.9 ton/ha, root 2.6 ton/ha, leaf 0.79 ton/ha, leaf area index 0.37, undergrowth of sedge 2.57 ton/ha, tree density 200/ha, relative basal area of trees 0.35 o/oo.
7) It was found that the biomass of small trees (4.5 cm>DBH>1 cm) and ground vegetation (4.5 cm <= DBH) was so unevenly distributed over the forest floor that a few hundred square meters of sample area would be needed for estimating them at a moderate level of statistical reliability.
8) The estimated biomass of the evergreen seasonal forest was compared with the data hitherto obtained in moist tropical forests of Cote d'Ivoire and Thailand. The forest of Cheko was found to have the biomass equivalent to other rain forests, but to be characterized by a specific DBH-tree height curve, a rather small leaf area index and a high value of leaf area/leaf weight ratio.
G-protein coupled receptors (GPCRs) comprise the largest superfamily of cell surface receptors and possess a signature motif of seven transmembrane helices. The endothelin B (ETB) receptor is a member of rhodopsin like GPCR family. It plays an important role in vasodilation and is found in the membranes of the endothelial cells enveloping blood vessels. Knowledge of the three-dimensional structure of G-protein coupled receptors in general would significantly add to our understanding of their molecular mechanisms and would be useful in the search for new specific drugs. However, three-dimensional structural analysis will require milligram quantities of pure and homogeneous protein. This dissertation is a study of the production, biochemical characterization and preliminary structural studies of the human ETB G-protein coupled receptor. The present work aimed at elucidating the structure and mechanistic details of function of the receptor by using a combination of X-ray crystallographic and NMR methods for collecting structural data. To obtain homogenous and monodisperse receptor protein preparation for structural and functional studies, we implemented the baculovirus expression system for the production of ETB receptor for the present work. The two step affinity purification ensured capture of full-length receptor. Silver stained SDS-PAGE of the purified receptor-ligand complex indicated greater than 90% protein purity. Based on previous reports, we used the high affinity ligand (endothelin -1) binding to the receptor for co-crystallization of receptor-ligand complex by locking the receptor in the activated conformation. As a prerequisite for 3D crystallization trials, the stability of the detergent solubilized receptor-ligand complex was assessed with respect to pH, temperature and time. Receptor-ligand complex did not show any degradation and aggregation over 6 days at 4°C and 18°C. Interestingly, change of pH suggested that receptor-ligand complex is unstable at lower pH due to possible charge induced conformational changes. In our work, we introduced the idea of using fluorophore labeled ligand for simple visual recognition of the receptor-ligand complex during purification and crystallization. On the other hand, we alternatively used biotinylated endothelin-1 to produce an adequate amount of ligand bound receptor complex, thus ensuring homogeneity of the purified complex for use in structural studies. Thus far, preliminary crystals have been obtained for both the unlabelled ET-1 and fluorophore labeled ET-1 complexed with ETB receptor. Moreover, we performed the systematic investigation of the protein/peptide binding partner for the receptor-ligand complex with the chief aims of stabilizing structure and increasing the possibilities of 3D-crystal contacts. Thus subsequent to formation of receptor-ligand complex, the additional in vitro formation of a ternary arrestin-receptor-ligand complex was also attempted for use in structural studies. We successfully demonstrated that arrestin mutant (R169E) forms a tight complex with ETB receptor regardless of its phosphorylation state. A second approach to get insight into the ETB receptor ligand binding site relied on the use of spin isotope labeled ET-1 ligand peptide by employing solid state MAS NMR method. Preliminary data provided compelling evidence that the C-terminal region of the peptide is immobilized in an ordered environment and presumably bound to the receptor. This indicates that the approach is feasible, although there are difficulties in sample preparation for further spectral measurements and data collection which are currently being discussed in ongoing investigations. At this point of our research work, we initiated a collaborative effort to obtain high yields of pure, active receptor without post translational modifications, from an E. coli cell lysate based in vitro expression system. We successfully optimized the production of homogenous and monodisperse endothelin B receptor in mg amounts. Thus this could potentially provide an alternative source of high quality receptor production in large quantities for immediate crystallization trials. Thus we hope that the results from these investigations can be applied in a more general sense to the production and crystallization of other G protein-coupled receptors.
The heat stress response is characterized by the presence of heat stress transcription factors (Hsfs) which mediate transcription of heat stress genes. In tomato (Lycopersicon peruvianum) cell cultures the simultaneous expression of four Hsfs, which are either constitutively (HsfA1 and HsfA3) or heat-stress inducible (HsfA2 and HsfB1) expressed, results in a complex network with dynamically changing cellular levels, intracellular localization and functional interactions. In order to examine the relevance of their multiplicity as well as to get more insights into the complexity of the plant heat stress response, the individual tomato Hsfs were investigated with respect to their protein interactions in vitro and in vivo. To this aim, I used pull-down assays as well as yeast assays to study the following aspects: 1. Oligomeric state of Hsfs: the results show that all class A Hsfs (HsfA1, HsfA2 and HsfA3) are trimeric proteins and interact with each other via the oligomerization (HR-A/B) domain. The similarity of their HRA/B regions allows formation of homo- and heterooligomeric complexes between all class A Hsfs. This special property was investigated by mutational studies with HsfA2 indicating that the linker and the HR-B regions are the minimal part required for Hsf/Hsf interactions. The conserved hydrophobic amino acid residues of the HR-B region are most important whereas the amino acid residues of the linker may provide higher flexibility to the HR-B region. Another investigated factor was HsfB1. HsfB1 is a member of class B Hsfs, which are characterized by an oligomerization domain without the 21 amino acid residues linker inserted between the HR-A and HR-B regions. It has a low activator potential and exists exclusively as dimer. HsfB1 can not physically interact with class A Hsfs. However, HsfB1 and HsfA1, binding to adjacent HSE sites, are assumed to cause strong synergistic effects in gene activation. 2. Potential HsfB1 interacting proteins: we searched for HsfB1 interacting proteins by using recombinant His-tagged proteins with HsfB1 as baits in pull-down assays. Histones H2A, H2B and H4 were identified by means of Peptide Mass Finger Printing and N-terminal sequencing analyses. The three histones represent the major proteins in tomato whole cell extracts retrieved by HsfB1. 3. HsfA2/small heat stress proteins (sHsps) interaction: pull-down and yeast two-hybrid assays were used to study the specific interaction of HsfA2 with tomato class II sHsp. This interaction occurs via the oligomerization domain of HsfA2. Other members of the plant Hsp20 family, including class I sHsp, do not interact with HsfA2. Heterooligomers of HsfA2 with class II sHsp may represent precursor forms of the plant higher molecular weight cytoplasmic complexes of heat stress granules, which form during heat stress. The findings presented in this thesis are a contribution to support the concept of a Hsfs network via protein-protein interactions. These data, together with information obtained from other studies, are used to propose a tentative model of the complex Hsfs network controlling the plant heat stress response.
Natrium/Protonen-Austauscher sind integrale Proteine biologischer Membranen und aufgrund ihrer funktionalen Abhängigkeit von einem elektrochemischen Gradienten der Klasse der Sekundärtransporter zugeordnet. Sie spielen eine essentielle Rolle sowohl in der Adaption von Bakterien an eine saline, alkalische Umgebung, als auch in der Regulation des intrazellulären pH- und Natriumhaushalts in Eukaryonten. Aufgrund der medizinischen Relevanz, unter anderem im Rahmen in der Behandlung des Herzinfarkts, besteht großes Interesse an der Struktur und den biochemischen Charakteristika des im Menschen ubiquitär vorkommenden Natrium/Protonen-Austauschers NHE1. Die heterologe und funktional aktive Produktion eukaryontischer Membranproteine stellt jedoch immer noch eine enorme Herausforderung dar, bei der sich das auf dem Semliki Forest Virus basierende Expressionssystem als gut geeignet erwiesen hat. Da die Überexpression von NHE1 mittels verschiedener eukaryontischer Expressionssysteme bisher kein kristallisationsfähiges Material liefern konnte, sollte in dieser Arbeit die heterologe Gewinnung von NHE1 mit dem Semliki Forest Virus Expressionssystem ermöglicht werden. Das Semliki Forest Virus Expressionssystem wurde auf Basis eines Vektorkonstrukts mit GFP zur späteren Übertragung der Parameter auf die Produktion von NHE1 etabliert. Konstrukte von NHE1 mit N- und C-terminalem Affinitäts-Tag wurden erfolgreich kloniert und zur Infektion von BHK-21 Zellkulturen eingesetzt. Dabei konnte beobachtet werden, dass der N-Terminus abgespalten wird und wahrscheinlich als Signalpeptid zum Einbau in die Membran dient. Das Protein wurde im Endoplasmatischen Retikulum lokalisiert, wo die Glykosylierung zum Transport in die Plasmamembran unterbleibt, was auf eine Interferenz mit der Virusinfektion zurückgeführt wurde. Eine Infektion der Zellen mit dem Semliki Forest Virus hat neben einem bereits bekannten massiven Anstieg des intrazellulären Natriumgehalts eine starke Alkalinisierung des Zytoplasma zur Folge. Ähnliches ist bisher über die Infektion von Zellen mit dem Poliovirus bekannt und stellt dort ein Schlüsselelement in der Sicherstellung der viralen Replikation dar, was auch für das Semliki Forest Virus zu gelten scheint. Die Expression von NHE1 konnte im 8 Liter-Maßstab optimiert und sowohl die Präparation als auch die Solubilisierung mit verschiedenen Detergenzien erfolgreich eingeführt werden. NHE1 erfährt jedoch bereits in vivo einen erheblichen proteolytischen Abbau, der sich während der Membranpräparation und Aufreinigung fortsetzt und zu einer Fragmentierung führt, die trotz des Einsatzes unterschiedlicher Kultivierungszeiten, Detergenzien, Additive oder Proteaseinhibitoren in vivo als auch in vitro nicht in einem Maße reduziert werden konnte, welches zur Gewinnung von kristallisationsfähigem Material erforderlich gewesen wäre. Es muss empfohlen werden einen in vivo Ansatz zu etablieren, um die proteolytische Degradation zu unterdrücken. Da die Virusreplikation nicht erforderlich ist, wäre Bafilomycin als Inhibitor der V-Typ ATPase geeignet, um die intrazelluläre Alkalinisierung und somit wahrscheinlich den Abbau von NHE1 zu verhindern. Ebenso erscheint der Einsatz von MG-132 zur spezifischen Inhibierung des Proteasoms Erfolg versprechend, was aber wegen hoher Kosten praktisch kaum in Frage kommt. Da man trotz individuell gelagerter Unterschiede zwischen den einzelnen Natrium/Protonen-Austauschern von einem ähnlichen Prinzip in Regulation und Transport ausgeht, wurden Strukturuntersuchungen mit Hilfe der Kryo-Elektronenmikroskopie am bakteriellen Natrium/Protonen-Antiporter NhaA aus Escherichia coli durchgeführt, um die strukturelle Basis der pH-Wahrnehmung und die Translokation von Natrium in das Periplasma besser zu verstehen. Die vorliegende Röntgen- und EM-Struktur repräsentieren den inaktiven Zustand, weshalb der eigentliche Ablauf des Transportvorgangs bisher biochemisch herzuleiten war, da bislang keine Kristalle im aktiven Zustand gezüchtet werden konnten. Durch die in situ Inkubation von 2D-Kristallen konnten aktive Zustände des Proteins direkt auf dem EM-Netz induziert und kryo-elektronenmikroskopisch festgehalten werden. Einzelne Datensätzen wiesen Reflexe bis zu 5 Å auf. Aus den angefertigten Projektionsdichte- und Differenzkarten ergaben sich pH- und Natrium-abhängige Konformationsänderungen. Die Röntgenstruktur wurde mit Hilfe des Molekularen Ersatzes in die EM-Struktur eingepasst und diente der Zuordnung und Interpretation der beobachteten Zustände als Basis. Die pH-abhängige Konformationsänderung wurde einem mit der funktional wichtigen Helix 9 assoziierten Bereich zugeordnet, welcher durch die Röntgenstruktur nicht definiert ist und wahrscheinlich die fehlenden Aminosäuren des regulatorisch relevanten N-Terminus enthält. Die beobachtete Konformationsänderung stellt das Entstehen einer besser geordneten Struktur dar und geht mit der pH-regulierten Aktivierung von NhaA zwischen pH 6 und 7 einher, weshalb dieser Bereich des Proteins zumindest als Bestandteil des sogenannten pH-Sensors betrachtet werden kann. Nach der vollständigen Aktivierung durch den pH-Wert, welche der folgenden Natrium-abhängigen Konformationsänderung vorauslaufen muss, konnte beobachtet werden, dass die Präsenz von Natrium im Rahmen der Ionentranslokation eine Bewegung des periplasmatischen Teils von Helix 4 induziert. Es wäre interessant, eine tiefergehende und genauere Charakterisierung der beobachteten Konformationsänderungen durch die Erstellung einer dreidimensionalen EM-Dichtekarte zu ermöglichen. Des Weiteren hat die eingehendere Untersuchung des röntgenkristallographischen Monomers nach der Einpassung in das physiologisch vorliegende Dimer der EM-Struktur sowohl eine für Membranproteine neuartige „Joint β-Sheet“ Dimerisierungsdomäne im Periplasma, als auch eine Verzahnung von Helix 7 und 9 an der Monomer-Monomer-Grenze aufgezeigt. Diesen Charakteristika kommt wahrscheinlich eine tragende Rolle in der Dimerisierung von NhaA zu, was durch weitere Untersuchungen im Rahmen einer Mutagenesestudie unter Einbeziehung der periplasmatischen β-Haarnadelstrukturen überprüft werden sollte.
Im Rahmen der vorliegenden Arbeit wurden verschiedene proteomanalytische Methoden untersucht und evaluiert, die, basierend auf der Verwendung 2D-gelelektrophoretischer, 2D-chromatographischer und massenspektrometrischer Techniken, die differentielle, quantitative Proteinanalyse zweier unterschiedlicher muriner Fibroblasten-Zelllinien ermöglichen. Hierfür wurden zunächst unterschiedliche Methoden für die 2D-elektrophoretische Proteinauftrennung analysiert. Im Hinblick auf eine größtmögliche Auflösung und Gel-zu- Gel-Reproduzierbarkeit wurden innerhalb der ersten Dimension (IEF) die Lademethode, die Fokussierungszeiten und der Reduktionsschritt (DTT oder HED) optimiert. Desweiteren wurde eine auf isoelektrischer Fokussierung basierende Vorfraktionierungsmethode auf ihre Anwendbarkeit bei einer quantitativen Proteomanalyse getestet. Für die Proteingelfärbung wurde unter anderem ein selbst synthetisierter Fluoreszenzfarbstoff eingesetzt, der hinsichtlich Färbesensitivität und MS-Kompatibilität mit etablierten Protokollen verglichen wurde. Eine Doppelfärbungs-Methode von Proteingelen (Silberfärbung nach Fluoreszenzfärbung) wurde auf ihre MS-Kompatibilität nach tryptischen Verdau untersucht. Desweiteren wurden manuelle und automatisierte Verdauprotokolle für eine möglichst hohe Peptide Recovery optimiert. Die zunächst durch die Anwendung von klassischen Färbe- und Quantifizierungsmethoden nach 2DE gewonnenen Ergebnisse wurden mit neueren Labelling-Methoden zur relativen Proteinquantifizierung verglichen. Dabei kamen zwei unterschiedliche Multiplexing-Verfahren zum Einsatz, die sich in der Proteinquantifizierung grundlegend unterscheiden (DIGE: gelbasierte Proteinquantifizierung; iTRAQ: LC-MS/MS basierte Peptidquantifizierung). Die für diese beiden Methoden bestehenden Protokolle wurden für die Anwendbarkeit auf die Fibroblasten-Proteinextrakte angepasst. Es konnte gezeigt werden, daß diese beiden Labelling-Methoden in Bezug auf Reproduzierbarkeit und quantitativer Aussagekraft dem klassischen 2DE-Experiment (Proteinfärbung nach der Auftrennung auf einzelnen Gelen) überlegen sind. Die statistische Absicherung der analysierten relativen Quantitätsunterschiede verbesserte sich durch die zusätzliche Anwendung der beiden neuen Labelling-Methoden erheblich. Dabei stützt sich die Signifikanz der quantitativen Bestimmung sowohl auf die große statistische Sicherheit, die innerhalb dieser beiden Multiplexing-Methoden erreicht wird, als auch auf die Wiederholbarkeit in unterschiedlichen Experimenten (21 Proteine wurden in unterschiedlichen Ansätzen bestätigt). Die beiden Labelling-Methoden DIGE und iTRAQ unterscheiden sich außer in der Quantifizierungsstrategie auch grundsätzlich in dem Ansatz der Auftrennung (DIGE: Proteine, 2DElektrophorese; iTRAQ: Peptide, 2D-Flüssigchromatographie). Damit besitzen sie unterschiedliche Limitierungen in Bezug auf die physiko-chemischen Eigenschaften der Peptide/Proteine, die mit der jeweiligen Methode aufgetrennt werden können. Der daraus resultierende komplementäre Charakter beider Methoden konnte anhand mehrerer Proteine verdeutlicht werden. Durch die relative Quantifizierung konnten insgesamt 30 Proteine identifiziert werden, die aufgrund der An- oder Abwesenheit des MLL-Proteins in den beiden murinen Zelllinien differentiell reguliert sind. Die alleine schon durch die unterschiedliche Morphologie der untersuchten murinen Fibroblasten vermutete Deregulation von Struktur- und Stressproteinen (Actin, HSP27, HSP70) konnte bestätigt werden. Weitere Expressionsunterschiede zwischen Mll-/-- und Mll+/+-Fibroblasten zeigten sich vor allem bei Proteinen, die funktionell der Gruppe RNA-prozessierender Proteine (Polyadenylate Binding Protein, PTB-associated Splicing Factor, hnRNPs) zugeordnet werden können. Ein Vergleich der quantitativen Proteomdaten dieser Arbeit mit den mRNA-Expressionsprofilen der gleichen Zellen zeigt nur eine sehr geringe Korrelation bezüglich der Regulationen einzelner Gene/Proteine. Die meisten der bisherigen Studien, die eine Untersuchung des mRNA/Protein-Verhältnisses zum Gegenstand haben, bestätigen das Fehlen einer Korrelation. Diese Tatsache unterstreicht die Wichtigkeit der Kombination genomischer und proteinanalytischer Daten zur Aufklärung zellulärer molekularer Prozesse.
The mTOR kinase inhibitor rapamycin (sirolimus) is a drug with potent immunosuppressive and antiproliferative properties. We found that rapamycin induces the TGF/Smad signaling cascade in rat mesangial cells (MC) as depicted by the nuclear translocation of phospho-Smads 2, -3 and Smad-4, respectively. Concomitantly rapamycin increases the nuclear DNA binding of receptor (R)- and co-Smad proteins to a cognate Smad-binding element (SBE) which in turn causes an increase in profibrotic gene expression as exemplified by the connective tissue growth factor (CTGF) and plasminogen activator inhibitor 1 (PAI-1). Using small interfering (si)RNA we demonstrate that Smad 2/3 activation by rapamycin depends on its endogenous receptor FK-binding protein 12 (FKBP12). Mechanistically, Smad induction by rapamycin is initiated by an increase in active TGF1 as shown by ELISA and by the inhibitory effects of a neutralizing TGF antibody. Using an activin receptor-like kinase (ALK)-5 inhibitor and by siRNA against the TGF type II receptor TGF-RII) we furthermore demonstrate a functional involvement of both types of TGF receptors. However, rapamycin did not compete with TGFfor TGF-receptor binding as found in radioligand-binding assay. Besides SB203580, a specific inhibitor of the p38 MAPK, the reactive oxygen species (ROS) scavenger N-acetyl-cysteine (NAC) and a cell-permeable superoxide dismutase (SOD) mimetic strongly abrogated the stimulatory effects of rapamycin on Smad 2 and 3 phosphorylation. Furthermore, the rapid increase in Dichlorofluorescein (DCF) formation implies that rapamycin mainly acts through ROS. In conclusion, activation of the profibrotic TGFSmad signaling cascade accompanies the immunosuppressive and antiproliferative actions of rapamycin. Life-attenuated measles virus (MV) vaccines have revealed their capacity to routinely induce life-long immunity against MV after just a single or two low-dose injections. Moreover, MV vaccines have been shown to be extensively safe and well tolerated, in general. Thus, MV is a prime candidate for a recombinant vaccine platform to protect also against other pathogens after vaccination. For this purpose, foreign genes can be inserted into additional transcription units (ATU) in recombinant MV genomes so that the encoded foreign proteins are co-expressed with MV proteins in infected cells. These so-called bivalent MV should protect against infection by MV or the pathogen, which the encoded foreign protein had been derived from. Bivalent MVs have already been shown to be effective vaccines against e.g. dengue virus or hepatitis B virus infections by inducing humoral and sometimes also cellular immune responses. In most of these studies, soluble or soluble versions of the pathogens' antigens were used for generation of bivalent MVs.
We hypothesized that the form of the antigen expressed by bivalent MVs is crucial for the potency and constitution of the induced immune responses. Therefore, three different forms of an antigen expressed by bivalent MVs were analyzed, here. The model antigen chosen for this purpose has been the envelope protein (Env) of SIVsmmPBj1.9. In its natural mature form, Env is composed of the surface unit gp120 and the transmembrane unit gp41, which stay non-covalently linked after proteolytic processing of the common precursor protein gp160. However, gp120 can be shed by infected cells or virus particles. Therefore, natural gp160 antigen was used as shedding form. Furthermore, stabilized covalently-linked gp160 variants and soluble gp140 variants were used in this thesis. These different antigen forms were inserted either behind the P or behind the H expression cassettes into the MV genome. The respective bivalent MVs were rescued and characterized. Expression of SIVsmmPBj1.9 Env variants by the bivalent MVs was confirmed by immuno blot and in situ immunoperoxidase assays. Replication curves of bivalent MV showed that growth of MVs expressing the different Env variants was slightly delayed by approximately 24 h compared to control viruses.
For immunization of transgenic, MV-susceptible IFNAR-/--CD46Ge mice, which are the current standard to analyze MV vaccines in a small animal model, an optimal dose of 1x105 TCID50 was determined. For the evaluation of humoral immune responses in transgenic mice, two ELISA systems for the detection of total α-MV and α-SIV antibodies and neutralization assays for detection of neutralizing antibodies against MV and SIV in sera of immunized mice were established. Mice immunized with any of the bivalent MVs showed significant humoral immune responses against MV comparable to those elicited by the parental MV vaccine strain without further genetic modifications. Mice immunized with MVvac2-gp140(P) expressing the soluble gp140 variant revealed highest α-SIV titers with a maximal OD of up to 0.4. Second highest levels of α-SIV antibodies were detected in mice that were immunized with the shedding variants or soluble Env in other positions. MVs expressing the stabilized variants induced only very low α-SIV antibody titers. Neutralizing antibodies directed against SIV could be detected in sera of mice immunized with MVs expressing the soluble or shedding variants, but not in sera of mice immunized with MVs expressing the stabilized variants. In sera of control mice immunized with PBS no antibodies could be detected, as expected. Thus, soluble and shedding antigens induced humoral immune responses, whereas stabilized antigens induced only weak humoral immune responses but no neutralizing antibodies. Analysis of cellular immune responses is still ongoing.
Besides Env, further SIV antigens could be tested for their potency to induce humoral as well as cellular immune responses.
Besides being used as a vaccine platform, recombinant MVs are evaluated as future agent for cancer therapy due to their significant inherent tumor-lytic, so-called oncolytic activity. Currently, the anti-tumoral activity of MV is analyzed in clinical phase I trials. MV strains with high fusion activity are used as oncolytic agents. The fusion protein F of MV strain NSe is highly fusogenic, in contrast to e.g. F of MVwt323, a clone of the pathogenic strain IC-B. Sequence analysis of these two proteins identified one coding nucleotide difference at aa 94 in the F2 domain: a valine (V) in FNSe and a methionine (M) in Fwt323. To evaluate impact of this difference, residues at aa 94 were exchanged. After transient-transfection of MV F and H expression plasmids in receptor-positive cells, V94 in the F2 subunit of FNSe or Fwt323 led to about 6-fold higher fusion activity compared to F proteins with M94. The co-expressed H protein (HNSe or Hwt323) did not influence fusion activity, indicating that the receptor (CD46 or SLAM) bound by H does not quantitatively affect the F proteins' activation. Analysis of F and H showed that formation and transport of MV glycoprotein complexes are not altered by substitution in aa 94 of FNSe or Fwt323.
Furthermore, recombinant MVNSe, MVNSe-F-M94, MVwt323, or MVwt323-F-V94 were rescued. Viral replication revealed slightly higher titers for recombinant MVs expressing M94 in F after 96 h of replication, compared to MVs expressing V94. MVs expressing V94 in F2 showed 2.5-fold higher fusion activity on CD46- and SLAM-positive Vero-hSLAM cells and 2-fold higher fusion activity on B95a cells expressing only SLAM compared to MVs expressing F with M94. Fusion activity of recombinant MVs can thus be modulated by substituting a single aa. V94 in the F protein results in highly fusion active MVs with possibly increased direct cytotoxicity in infected tumors, whereas M94 in F could be associated with decreased fusion activity for therapies, where higher virus titers are required.
Reggie-1 (flotillin-2) and reggie-2 (flotillin-1) are membrane microdomain proteins which are associated with the membrane by means of acylation. They influence different cellular signaling processes, such as neuronal, T-cell and insulin signaling. Upon stimulation of the EGF receptor, reggie-1 becomes phosphorylated and undergoes tyrosine 163 dependent translocation from the plasma membrane to endosomal compartments. In addition, reggie-1 was shown to influence actindependent processes. Reggie-2 has been demonstrated to affect caveolin- and clathrin-independent endocytosis. Both proteins form homo- and hetero-oligomers, but the function of these oligomers has remained elusive. Moreover, it has not been clarified if functions of reggie-1 are also influenced by reggie-2 and vice versa. The first aim of the study was to further investigate the interplay and the heterooligomerization of reggie proteins and their functional effects. Both reggie proteins were individually depleted by means of siRNA. In different siRNA systems and various cell lines, reggie-1 depleted cells showed reduced protein amounts of reggie-1 and reggie-2, but reggie-2 knock down cells still expressed reggie-1 protein. The decrease of reggie-2 in reggie-1 depleted cells was only detected at protein but not at mRNA level. Furthermore, reggie-2 expression could be rescued by expression of siRNA resistant wild type reggie-1-EGFP constructs, but not by the soluble myristoylation mutant G2A. This mutant was also not able to associate with endogenous reggie-1 or reggie-2, which demonstrates that membrane association of reggie-1 is necessary for hetero-oligomerization. In addition, fluorescence microscopy studies and membrane fractionations showed that correct localization of overexpressed reggie-2 was dependent on co-overexpressed reggie-1. Thus, hetero-oligomerization is crucial for membrane association of reggie-2 and for its protein stability or protein expression. Moreover, the binding of reggie-2 to reggie-1 required tyrosine 163 of reggie-1 which was previously shown to be important for endosomal translocation of reggie-1. Since reggie-2 was implicated to function in clathrin- and caveolin-independent endocytosis pathways, the effect of reggie-2 depletion on reggie-1 endocytosis was investigated. Indeed, reggie-1 was dependent on reggie-2 for endosomal localization and EGF-induced endocytosis. By FRET-FLIM analysis it could be shown that reggie heterooligomers are dynamic in size or conformation upon EGF stimulation. Thus, it can be concluded that reggie proteins are interdependent in different aspects, such as protein stability or expression, membrane association and subcellular localization. In addition, these results demonstrate that the hetero-oligomers are dynamic and reggie proteins influence each other in terms of function. A further aim was the characterization of reggie-1 and reggie-2 function in actindependent processes, where so far only reggie-1 was known to play a role. Depletion of either of the proteins reduced cell migration, cell spreading and the number of focal adhesions in steady state cells. Thus, also reggie-2 affects actin-dependent processes. Further investigation of the focal adhesions during cell spreading revealed that depletion of reggie-1 displayed different effects as compared to reggie-2 knock down. Reggie-1 depleted cells had elongated cell-matrix-adhesions and showed reduced activation of FAK and ERK2. On the other hand, depletion of reggie-2 resulted in a restricted localization of focal adhesion at the periphery of the cell and decreased ERK2 phosphorylation, but it did not affect FAK autophosphorylation. Hence, reggie proteins influence the regulation of cell-matrix-adhesions differently. A link between reggie proteins and focal adhesions is the actin cross-linking protein -actinin. The interaction of -actinin with reggie-1 could be verified by means of co-immunoprecipitations and FRET-FLIM analysis. Reggie-1 binds -actinin especially in membrane ruffles and in other locations where actin remodeling takes place. Moreover, -actinin showed a different localization pattern during cell spreading in reggie-1 depleted cells, as compared to the control cells. These results provide further insights into the function of both reggie proteins. Their interplay and hetero-oligomerization was shown to be crucial for their role in endocytosis. In addition, both reggie proteins influence actin-dependent processes and differentially affect focal adhesion regulation.
In der vorliegenden Dissertation stand die Aufklärung der Funktion und Regulation von p21 in der Mitose im Mittelpunkt. p21 ist als Cdk-Inhibitor und Schlüsselregulator bekannt, der in viele fundamentale zelluläre Prozesse involviert ist: Zellzyklusregulation, Apoptose, Seneszenz, Zellmigration und Dynamik des Zytoskeletts, Transkription, Differenzierung sowie DNA-Reparatur, aber auch in die Umprogrammierung induzierter pluripotenter Stammzellen (Besson et al. 2008; Abbas und Dutta 2009; Jung et al. 2010).
Die unkontrollierte Proliferation von Zellen ist mit der Tumorgenese assoziiert und wird unter anderem durch die Fehlregulation von p21, aber auch durch die wichtigen mitotischen Kinasen Cdk1, im Komplex mit ihrer regulatorischen Untereinheit Cyclin B1, sowie Plk1 bedingt. Zudem ist das Fehlen von p21 oder die Fehllokalisation in das Zytoplasma mit einer schlechteren Prognose für den Patienten und Chemotherapie-Resistenz von Tumoren verbunden (Abukhdeir und Park 2008). Aufgrund der zunehmenden Inzidenz und Mortalität von Krebserkrankungen ist es daher von besonderem klinischem Interesse, die molekularen Ursachen für die Entstehung maligner Tumorerkrankungen aufzuklären. Bislang existieren kaum Studien über welche molekularen Mechanismen die Funktionen von p21, dem wichtigsten Cdk-Inhibitor, der zum Beispiel durch die Anwendung niedermolekularer Inhibitoren wie BI 2536, das sich bereits in klinischen Phase II Studien befindet (Strebhardt 2010), beeinflusst wird, während der Mitose reguliert werden.
In der vorliegenden Dissertation wurde daher die physiologische Rolle des Cdk-Inhibitors bzw. Regulators p21 während der Mitose untersucht und mit der Kinaseaktivität von Cdk1/Cyclin B1, wie auch Plk1 korreliert. Es konnte gezeigt werden, dass p21 während der Mitose stark exprimiert wird und dass mitotisches p21 in verschiedenen Krebszelllinien unabhängig von dem p53-Status in einer phosphorylierten Form vorkommt, welche mit der Aktivität von Cdk1 und weniger mit der von Cdk2 assoziiert ist. Durch Untersuchungen der isogenen HCT116-Zelllinien mit und ohne p21 wurde aufgezeigt, wie wichtig p21 für den ordnungsgemäßen Ablauf der Mitose ist. Ohne p21 sind sowohl die Anaphase wie auch die Zytokinese verlängert, die Zellen ordnen die Chromosomen fehlerhaft in der Metaphaseplatte an (congression Fehler), besitzen weitaus mehr lagging Chromosomen und fast 20 % der Zellen weisen im Versuchsverlauf Polyploidie auf. Durch den Verlust des Cdk-Regulators p21 kommt es zur Fehlregulation von Cdk1 und seiner Substrate (wie MCAK) und es treten die oben beschriebenen Probleme auf.
Weiterhin phosphoryliert Cdk1/Cyclin B1 p21 an Ser-130 in vitro und ex vivo in der frühen Phase der Mitose, der Prophase bzw. Prometaphase. Die nicht phosphorylierbare p21 Form S130A befindet sich hauptsächlich im Zellkern und führt zu vermehrtem Auftreten von congression Fehlern, während die S130D-Mutante, die die Phosphorylierung durch Cdk1 vortäuscht, schneller degradiert wird und zudem den Phänotyp der HCT116 p21-/- Zellen verstärkt. Zellen, die S130D exprimieren, benötigen mehr Zeit für das Durchlaufen der Mitose. Hier ist vor allem die Metaphase stark verlängert, aber auch Anaphase und Zytokinese. Dies führt zu congression Fehlern und zu Polyploidie. Diese Ergebnisse bestätigen, wie wichtig die zeitlich korrekte Phosphorylierung von p21 und die dadurch vermittelte Aktivierung von Cdk1/Cyclin B1 ist.
Darüber hinaus stabilisiert die Suppression von Plk1 das p21 Protein, was darauf hinweist, dass die Degradation von p21 während der Prometaphase von Plk1 kontrolliert wird. Dies wird von der Tatsache unterstützt, dass Ser-114, wie auch Ser116 von Plk1 in vitro phosphoryliert wird. Die Deregulation von p21 durch Plk1, SS114/116AA bzw. SS114/116DD induziert Chromosomenfehler, wodurch die molekularen Mechanismen, warum fehlreguliertes Plk1 die Tumorgenese fördert, hervorgehoben werden.
Nach Abschluss der bisherigen Untersuchungen steht fest, dass man sich von der starren Rolle von p21 als Tumorsuppressor und Akteur während der G1/S-Phase lösen muss. Der Cdk-Inhibitor p21 trägt entscheidend zur mitotischen Progression bei, vor allem bedingt durch die zeitlich ordnungsgemäße Inaktivierung bzw. Aktivierung von Cdk1/Cyclin B1, der Kinase, die wiederum zahlreiche für die Mitose essentielle Proteine reguliert. In Zukunft muss zum besseren Verständnis der Rolle von p21 in der Mitose die genaue Abfolge der Ereignisse unter Einbeziehung der Degradationsmechanismen eingehender untersucht werden.
In this thesis I have investigated the regulation of eicosanoid synthesizing-enzymes by cannabinoid receptor agonists. Rat renal mesangial cells were used as a model system. I could show that all three (CB1, CB2, and GPR55) cannabinoid receptors are expressed on the mRNA level in rat renal mesangial cells – but with differing expression profiles. The CB1 and GPR55 receptors are expressed in comparable amounts, whereas the CB2 receptor is considerably less expressed than the CB1 and the GPR55 receptors. Furthermore I could show that stimulation of renal mesangial cells with CB1 receptor agonists, such as R(+)MA or ACEA, increased IL-1β-induced cPLA2, sPLA2-IIa, and COX2 protein and mRNA expression which subsequently led to an enhanced IL-1β-induced PGE2 formation. Additionally, the IL-1β- induced sPLA2-IIa promoter activity was also increased by CB1 receptor stimulation. Besides the modulated expression of the eicosanoid synthesizing enzymes, I could show that CB1 agonists also led to an increase of IL-1β-induced iNOS expression and subsequent NO formation. In contrast, stimulation with CB2 selective agonists led to a decrease in IL-1β- induced sPLA2-IIa protein expression and PGE2 formation. Accordingly, the IL-1β-induced sPLA2-IIa promoter activity was also reduced by CB2 receptor agonists. IL-1β-induced iNOS expression and subsequent NO formation were not influenced by CB2 recptor activation. Matching the results I obtained with CB1 receptor agonists on IL-1β-induced PGE2 formation, I could observe an increased cPLA2 protein and mRNA expression with a subsequent increase in IL-1β-induced PGE2 formation by GPR55 stimulation. Stimulation with THC, an unselective CB agonist, increased the IL-1β-induced sPLA2-IIa protein expression and subsequently led to an enhanced IL-1β-induced PGE2 formation. Subjecting the cells to higher THC concentrations surprisingly led to a reduction of the IL-1b-induced sPLA2-IIa protein expression and PGE2 formation. A possible explanation may be the differential expression of the three CB receptors. At low concentrations THC may predominantly activate CB1 and GPR55 and with increasing concentration CB2 receptors may also be activated, slightly reversing the enhancing effect. Moreover, I could show that the CB1 receptor stimulation mediated phosphorylation and hence the activation of ERK1/2 MAPK. Additionally to ERK1/2, there was also a phosphorylation and activation of NFkB observed by CB1 receptor stimulation. In my thesis I could show for the first time that PPARα was activated by IL-1β in rMC. The IL-1β-induced PPARα promoter activity was completely inhibited by addition of the CB2 receptor agonist, JWH015. These findings were confirmed by inhibition of the IL-1β-induced PGE2 formation by a PPARα antagonist (MK-886). In summary, I could show that activation of CB1 receptors in our system led to a worsening of an inflammatory condition, whereas activation of the CB2 receptors led to the complete opposite; namely a reduction of the inflammatory response by reducing the sPLA2-IIa expression and PGE2 formation. GPR55 activation did not display any alteration of inflammatory conditions, since the classical inflammatory pathway was not influenced.
The generation of O2- by NADPH oxidaes was mainly attributed to immune cells that kill invading bacteria or cancer cells. But importantly, in the past several years, several homologs of the catalytic subunit gp91phox (Nox2) of the phagocytic NADPH oxidase have been identified in non-immune cells and tissues. Superoxide production derived from NADPH oxidaes has been shown to play a role not only in host defense but also in defined signaling cascades mediating growth and apoptosis. The aim of this work was to study the expression and the regulation of the”new” Nox isoforms in rat renal mesangial cells (MC). In particular the following results were achieved. 1) mRNA’s for both Nox1 and Nox4 were detected by RT-PCR. 2) Nox1 mRNA levels were increased upon exposure to basic fibroblast growth factor (bFGF), platelet-derived growth factor (PDGF) and fetal calf serum (FCS) in a time- and dose-dependent manner. Exposure of MC to bFGF and FCS increased also basal production of reactive oxygen species (ROS) by MC. By contrast, Nox4 mRNA levels were not significantly affected by bFGF treatment, but were markedly down-regulated by PDGF and FCS. 3) To study the regulation of Nox1 on the protein level, an anti-Nox1 antibody was generated and characterized using affinity chromatography. Up-regulation of Nox1 expression by growth factors was confirmed also on the protein level. 4) Based on the already known cDNA sequence for Nox1, the transcriptional start site was determined by the “gene RACE” technique. 2547 bp of the genomic sequence of the 5´-flanking region of the Nox1 gene were cloned and sequenced using the „Genome-Walking“ method. To study the regulation of Nox1 transcription functional Nox1 promoter/luciferase fusions were be established. MC were transiently transfected with different promoter/luciferase constructs and stimulated with growth factors. By measuring luciferase activity it was determined that growth factors induced the Nox1 transcription and that the Nox1 core promoter is sufficient for the activation. 5) By measurement of superoxide radicals and analysis of Nox1 mRNA expression by quantitative RT-PCR (TaqMan) as well as protein level by Western blotting it could be shown that treatment of MC with NO donors inhibited the expression of Nox1 in a time- and dose-dependent manner. Moreover, using activators and inhibitors of the soluble guanylyl cyclase (sGC) it could be shown, that the activation of sGC mediates the effect of NO on Nox1 expression. However, NO had no inhibitory effect on Nox1 promoter activity. Experiments with the inhibitor of transcription, actinomycin D, suggest that NO-mediated regulation of Nox1 is triggered probably via post-transcriptional mechanisms. Nox4 is regulated on the mRNA levels in a similar manner as Nox1. 6) To analyze the sub-cellular localization of the Nox isoforms, coding sequences for Nox1 and Nox4 were fused together with green fluorescent protein into the pEGFP-N1 demonstrated that both isoforms are localized predominantly in the plasma membrane, but also in the perinuclear region and cytoplasm. However, the localization of Nox1 in the plasma membrane was more pronounced. 7) In addition to Nox1 and Nox4, mRNA of the newly identified NOXA1 that is a homolog of the p67phox subunit of NADPH oxidase was detected in MC by RT-PCR.
NK cells are part of the innate immune system, and are important players in the body’s first defence line against virus-infected and malignantly transformed cells. While T cells recognize neoplastic cells in an MHC-restricted fashion, NK cells do not require prior sensitization and education about the target. In leukemia and lymphoma patients undergoing allogeneic hematopoietic stem cell transplantation not only T cells but also NK cells have been found to mediate potent graft-versus-tumor effects. Hence, autologous or donor-derived NK cells hold great promise for cancer immunotherapy. Since the generation of highly purified NK cell products for clinical applications is labor-intensive and time consuming, established human NK cell lines such as NK-92 are also being considered for clinical protocols. NK-92 cells display phenotypic and functional characteristics similar to activated primary NK cells. While NK-92 cells are highly cytotoxic towards malignant cells of hematologic origin, they do not affect healthy human tissues. NK-92 cells can be expanded under GMP-compliant conditions, and can therefore be provided in sufficient numbers with defined phenotypic characteristics for clinical applications.
