The title molecule, C34H28I4·4C6H6, has crystallographic 4 symmetry and crystallizes with four symmetry-related benzene solvent molecules. The phenyl group is eclipsed with one of the adamantane C—C bonds. The tetraphenyladamantane units and the benzene solvent molecules are connected by weak intermolecular phenyl–benzene C—H⋯π and benzene–benzene C—H⋯π interactions. In the crystal, molecules are linked along the c-axis direction via the iodophenyl groups by a combination of weak intermolecular I⋯I [3.944 (1) Å] and I⋯π(phenyl) [3.608 (6) and 3.692 (5) Å] interactions.
Die vorliegende Arbeit hat die Charakterisierung und Untersuchung des Stabilitätsverhaltens von
Parvulustat (PL), einem Homologen des α-Amylase-Inhibitors Tendamistat, zum Inhalt. Zur
weitreichenden Charakterisierung wurden verschiedene Proteinregionen des Parvulustats der CTerminus,
das hydrophobe Cluster, die Disulfidbrücken sowie die Proline auf ihren jeweiligen
Einfluss auf die Struktur und die thermodynamische Stabilität untersucht. In der vorliegenden
Zusammenfassung werden die Ergebnisse dieser Studien komprimiert präsentiert:
· Charakterisierung des Parvulustat-Wildtyps
Es galt vorweg herauszufinden, wie sich der Inhibitor unter nativen und denaturierten
Bedingungen verhält, um Rückschlüsse auf seine Merkmale und Strukturen zu ziehen. Die
erzielten Ausbeuten und die hohe Reinheit des isolierten Parvulustats erlaubten eine umfassende
Charakterisierung, einschließlich zahlreicher Kristallisationsexperimente, die vermuten lassen,
dass eine Kristallisation möglich sein sollte. Aufgrund der Tatsache, dass die Struktur des
Parvulustats bis 2009 unbekannt war, wurde die sekundäre Struktur mittels Circulardichroismus
und Fluoreszenzspektroskopie untersucht. Die Analyse des fernen UV-CD-Spektrums bei pH 7,0
und 25°C offenbarte eine „all-b-sheet“ Protein-Struktur. Mittels Fluoreszenzspektroskopie wurde
deutlich, dass die aromatischen Aminosäuren exponiert vorliegen. Um zunächst einen Einblick in
die Strukturveränderungen und die thermodynamische Stabilität zu erhalten, wurde der
temperaturinduzierte Entfaltungsübergang mittels CD-Spektroskopie verfolgt. Das Angleichen der
bei 230 nm gemessenen CD-Daten nach der linearen Extrapolations-Methode für eine
Zweizustands-Faltung ergab einen Tm-Wert von 82°C und D H(Tm) von 201,6 kJ/mol. Die
beträchtlichen Werte veranschaulichen die hohe Stabilität des Parvulustats. Eine aus den CDMessungen
bei 50° ergebende Übergangskurve zeigte, dass sich die Sekundärstruktur mit einem
Übergangsmittelpunkt bei 5,62 M GdnHCl kooperativ und reversibel entfaltet. Das Protein
entfaltet sogar infolge einer pH-Wert-Senkung bis auf pH 1 nicht vollständig, sondern es wechselt
direkt in einen Säure-Zustand („acid-state“). Dieser Zustand zeigt spektroskopisch die gleichen
Eigenschaften wie das native Protein, wobei die volle Inhibierungs-Aktivität nicht erhalten bleibt.
In sehr basischem Milieu bei pH 14 nimmt Parvulustat einen alkalisch denaturierten
Zwischenzustand IB an, der sich erheblich von dem GdnHCl-denaturierten, dem säurebehandelten
oder vom „molten globule“ Zustand unterscheidet. Allgemein behielt Parvulustat
über ein breites pH-Wert Spektrum (1,0-10,0) die native Struktur, bzw. eine „native like“ Struktur,
was erneut auf die enorme Stabilität des Proteins hindeutet. Die von Rehm et al. (Rehm et al.,

2009) aufgeworfene Hypothese des „induced fit“ Inhibierungsmechanismus des Parvulustats
konnte durch die in dieser Arbeit durchgeführten Experimenten bekräftigt werden. Mittels
Sekundärstrukturbestimmungen des Parvulustats unter Komplexbildung mit der a-Amylase
konnte eindeutig gezeigt werden, dass strukturelle Veränderungen am Inhibitor im Komplex
vorliegen. Durch zahlreiche Tests konnte festgestellt werden, dass die WRY-Region des
Parvulustats sich der Struktur der a-Amylase anpasst. Die Komplexierung des Parvulustats
bewirkte aber eine thermodynamische Destabilisierung der Inhibitor-Struktur.
· Einfluss des C-Terminus auf die Stabilität des Parvulustats
Um die Ursachen für die hohe Stabilität des Parvulustats auch im Vergleich zu Tendamistat (Tm:
79°C) zu finden, wurde der Einfluss des hoch flexiblen C-Terminus untersucht. Die Derivate mit
um zwei (PL-2AA), vier (PL-4AA) und sieben (PL-7AA) Aminosäuren verkürztem C-Terminus
wurden isoliert und analysiert. Die Entfaltungstemperaturen der verkürzten Derivate des
Parvulustats sinken mit abnehmender Zahl der Aminosäuren. Die Ergebnisse suggerieren, dass
der C-Terminus des Parvulustats eine entscheidende Rolle in der strukturellen Vollständigkeit des
Proteins während der thermischen Entfaltung spielt und damit auch in der Faltung (Tab.: 2.1). Der
Vergleich der Inhibitoraktivitäten der verkürzten Proteine mit dem nativen Parvulustat ergibt für
die Varianten PL-2AA und PL-4AA eine dem Wildtyp ähnliche Aktivität. Die Variante PL-7AA
weist eine leicht verringerte Aktivität auf.

Einfluss hydrophober Oberflächenclustern auf Stabilität und Faltung
des Parvulustats
Parvulustat besitzt in der Mitte des ersten b-Faltblatts, um die Position 22 liegend, einen
hydrophoben Oberflächencluster. Wie mit Hilfe von Substitutionsexperimenten in dieser Region
gezeigt werden konnte, ist die thermodynamische Stabilität des Parvulustats in hohem Maße von
der Bildung dieses kleinen aber wichtigen hydrophoben Kerns bestimmt. Demnach muss das b-
Faltblatt I und das b-Hairpin I eine entscheidende Rolle in der Faltung von Parvulustat spielen.
Position 22 ist in zweierlei Hinsicht für die Stabilität des Parvulustats wichtig: einerseits durch die
energetisch wichtigen Wasserstoffbrückenbindungen und zum zweiten steuert die Stelle zur
Formation des hydrophoben Kerns bei. Die Daten suggerieren, dass es auch eine
thermodynamische Kopplung zwischen dem hydrophoben Effekt und der Präsenz von
Wasserstoffbrückenbindungen geben könnte. Zumindest aus der Sicht der Stabilität ist es
eindeutig, dass interatomare Interaktionen wie Wasserstoffbrückenbindungen, van-der-Waalsund
Dipol-Dipol-Wechselwirkungen notwendig sind, um eine stabile Bildung von b-Faltblättern
in Parvulustat und seinen Derivaten zu verwirklichen.
· Der Effekt der Proline auf die Stabilität des Parvulustats
Der Effekt der Substitution des Prolins durch Alanin an verschiedenen Positionen des Parvulustats
wurde ebenfalls untersucht. Im Ganzen betrachtet führt auch die Mehrfachsubstitution von Prolin
zu keiner nennenswerten strukturellen Veränderung im Parvulustat. Die durchgeführten
thermischen Entfaltungsexperimente bestätigen diese Beobachtungen. Alle Einzelmutanten (P5A,
P42A, P48A, P72A) zeigten eine höhere thermische Stabilität als der Wildtyp (Abb. 2.1).

Die fast gleich bleibenden Tm-Werte bzw. die Erhöhung der Stabilität des Parvulustats sind durch
die strukturelle Fluktuationen zu erklären, denn die rigide XAS-Pro-Bindung wurde durch die
flexible XAS-Ala-Bindung ersetzt.
· Der Einfluss der Disulfidbindung auf die Stabilität des Parvulustats
Der Einfluss der zwei Disulfidbrücken des Parvulustats wurde bezüglich Aktivität, spektraler
Eigenschaften sowie Stabilität untersucht. Hierfür wurden 25 Inhibitorvarianten mittels gezielter
Mutagenese gewonnen, in denen jeweils zwei Cysteinreste, die im natürlich vorkommenden
Parvulustat Disulfidbrücken bilden, durch andere Aminosäuren ersetzt wurden. Die Ergebnisse
zeigten, dass die Faltung Parvulustats ein zwei Zustand Verhalten besitzt, das außer in
Tendamistat in keinem anderen disulfid-verknüpftem Protein gefunden wurde. Dieses Verhalten
wurde durch die Entfernung von Disulfidbrücken nicht beeinflusst. Wie durch die CD- und
fluoreszenzspektroskopischen Experimente belegt werden konnte, ist die native Struktur des
Parvulustats durch die Entfernung der C43-C70-Disulfidbindung tiefgreifend verändert worden.
Passend zu den Veränderungen der Struktur haben die Mutationen schwerwiegende
Destabilisierungseffekte auf das Protein verursacht, was auch an der Erniedrigung der Freien
Gibbs Energie der Denaturierung und der Tm-Werte zu erkennen ist. Die Untersuchung des
Einflusses der Aminosäure-Substitution in den Positionen 43 und 70 auf die thermodynamische
Stabilität des Parvulustats führt zum Ergebnis, dass die Hydrophobizität und Polarität des Restes
70 einen bedeutenden Effekt auf die Stabilität des Proteins besitzt. Die Betrachtung der
thermodynamischen Daten macht deutlich, dass der Beitrag der freien Energie zur Stabilisierung
nicht nur abhängig von der eingeführten Aminosäure ist, sondern zum Teil auch von dem
strukturellen Kontext abhängt. Die Daten zeigen, dass die Substitution des Alanins an der Stelle
70 durch Leucin oder Threonin die Struktur um 0,3 bzw. 1,7 kJ/mol stabilisieren. Zusätzliche
Beiträge zum Unterschied bezüglich des ΔG°-Werts zwischen den Varianten C43AC70L/T und
C43L/TC70A können sich auch durch die unterschiedliche lokale Umgebung, wie z.B. die
Seitenketten von benachbarten Aminosäuren um die zwei Mutationsstellen ergeben. Der Tm-Wert
des Wildtyp-Proteins beträgt 82°C. Die Vergleiche dieser Größe mit der von der stabilsten
Cystein-defizitären Doppelmutante C43AC70T (47,3°C) ergibt eine Differenz von 34,7°C. Dieses
Ergebnis untermauert, dass die Disulfidbindung 2 eine extrem wichtige strukturelle Komponente
in der ungewöhnlich hohen Stabilität des Parvulustats darstellt.
Das Ziel der Arbeit war es dennoch die Daten der Stabilitäten einzelner Disulfidmutanten zu
sammeln und zu erfassen, um dadurch allgemeine Grundregeln für eine rationale Gestaltung der
Elimination beider Disulfidbrücken im Sinne einer vorhersagbaren Auswirkung auf die
Proteinstabilität zu bekommen.

Der Versuch ein disulfidfreies Protein in großen Mengen aus S. lividans zu isolieren, stellte sich
aber als extrem schwierig dar. Nach der Änderung des Stamms des Wirtsorganismus
(Streptomyces lividans TK23), Erhöhung der Qualität der Protoplasten und der Expressions-
Bedingungen (19°C, 150 rpm) konnten auf dem SDS-Gel stärkere Banden des Derivats
C9AC25TC43AC70T-4AA beobachtet werden. Die Expression der Mutante
C9AC25TC43AC70L-4AA konnte hingegen nicht nachgewiesen werden. Die anschließende
Reinigung des Derivats C9AC25TC43AC70T-4AA des Parvulustats mittels Gelfiltration und RPHPLC
bzw. Isolierung des Proteins aus dem SDS-Gel brachte das erwünschte Produkt. Dieses
konnte mit der MALDI-Massenspektroskopie eindeutig nachgewiesen werden. Das aktive
Cystein-freie Derivat C9AC25TC43AC70T-4AA konnte auch auf dem a-Amylase Plattentest
nachgewiesen werden. Diese Ergebnisse zeigen, dass eine Expression und der Nachweis einer
cystein-freien Variante möglich sind, dennoch haben die Ausbeuten dieses Proteins für
weiterreichende Analytik nicht ausgereicht. Demnach sind die Disulfidbrücken für die Stabilität
und Struktur des Parvulustats von enormer Bedeutung dennoch sind sie nicht zwingend
erforderlich für die Aktivität und die Expression in S. lividans.
The display of foreign polypeptides and proteins on the surface of viruses or cells provides an important tool for the engineering of biomolecules and the analysis of their interactions with binding partners. The most extensively used display platform is the coat protein of the filamentous bacteriophage (Smith, 1985). Phage display libraries have often been selected for polypeptides, e.g. single chain (sc) antibodies that bind to a protein of interest, but in vivo selection could only be demonstrated for peptides so far. An alternative display platform is the retrovirus murine leukemia virus (MLV). Here, polypeptides are displayed at the N-terminus of the viral envelope glycoprotein. Proof of principle for this platform was demonstrated for protease substrate libraries, which can be selected through coupling proteolytic activation with viral infectivity (Buchholz et al., 1998). Selection of the library CX4A on living cells resulted in viruses with more than three orders of magnitude improved spreading efficiency through tumor cells (Hartl et al., 2005). Also scAb libraries have recently been displayed and selected using retroviruses (Urban et al., 2005). The library scFvlibxMo displays the repertoire of phage display preselected sc antibodies for laminin-1 binding. The retrovirus based selection process resulted in laminin-specific sc antibodies with improved expression levels in mammalian cells.
This thesis describes the in vivo (i.e. in mouse tumor models) selection of the C-X4-A and scFvlibxMo for tumor homing upon systemic delivery.
For selection of the protease substrate library C-X4-A a subcutaneous tumor was induced in SCID mice followed by three systemic injections of the library. The selection process was monitored over a period of 34 days. After the incubation period mice were sacrificed and virus load in organs and tumor determined. PCR analysis after 34 days showed that virus from the library had preferentially infected the tumor. Sequence analysis showed the selection of protease substrates with the most prominent one with a frequency of over 65%. The four most prominent protease substrate variants where reconstituted into the original viral backbone for further investigation (C-SK-A, C-HI-A, C-HM-A and C-HS-A). Interestingly, these viruses exhibited a reduced spreading capacity in vitro on HT1080 cells as compared to the C-AK-A virus, which had previously been selected on HT1080 cells. When assayed for tumor homing, however, viruses C-HI-A and C-HS-A had clearly improved in comparison to C-AK-A. Tumor tissue had been infected at rates of over 55% while virus load of extratumoral organs was very low (infection rates <0.7 for C-HS-A and <0.02 for C-HI-A). Tumor targeting capacity had thus been improved over 10-fold by the in vivo selection of the C-X4-A library.
The experimental set up for the in vivo selection of the scFvlibxMo library was performed according to that of the C-X4-A library. Fingerprint analysis of the selected viruses that infected tumor tissue resulted in the identification of seven antibody variants showing unique CDR3 sequences. Two prominent clones (M49T-A and M49T-B) were cloned back into the MoMLV genome for further analysis of the reconstituted viruses. While variant B bound laminin-1 efficiently, variant A was unable to do so, although it was selected at highest frequency (76%). Both reconstituted viruses were equally well infectious and spread through HT1080rec1 cells at a similar efficiency as MoMLV. In an in vivo competition experiment the selected viruses clearly out-competed a laminin-1 binding reference virus L36xMo for tumor homing. To understand the molecular driving forces behind the in vivo selection process the epitope of the selected scFv M49T-A was identified using a phage peptide library approach. In silico analysis led to the identification of a small group of possible antigens, including tenascin, fibronectin and collagen.
The data described in this thesis demonstrate that the retrovirus display platform is capable of allowing the in vivo selection of protease substrates and scFvs. Notably, the replication competence of the system introduced an additional level of complexity to the library. The performed in vivo selections significantly enhanced tumor tropism. Selective infection of tumor cells combined with transfer of anti-tumoral genes is an attractive strategy for cancer therapy being in focus of current research. The viruses selected in this thesis build prime candidates for targeted retrovirus based tumor therapy.
Sucrose- and H+-dependent charge movements associated with the gating of sucrose transporter ZmSUT1

Background: In contrast to man the majority of higher plants use sucrose as mobile carbohydrate. Accordingly proton-driven sucrose transporters are crucial for cell-to-cell and long-distance distribution within the plant body. Generally very negative plant membrane potentials and the ability to accumulate sucrose quantities of more than 1 M document that plants must have evolved transporters with unique structural and functional features.
Methodology/Principal Findings: To unravel the functional properties of one specific high capacity plasma membrane sucrose transporter in detail, we expressed the sucrose/H+ co-transporter from maize ZmSUT1 in Xenopus oocytes. Application of sucrose in an acidic pH environment elicited inward proton currents. Interestingly the sucrose-dependent H+ transport was associated with a decrease in membrane capacitance (Cm). In addition to sucrose Cm was modulated by the membrane potential and external protons. In order to explore the molecular mechanism underlying these Cm changes, presteady-state currents (Ipre) of ZmSUT1 transport were analyzed. Decay of Ipre could be best fitted by double exponentials. When plotted against the voltage the charge Q, associated to Ipre, was dependent on sucrose and protons. The mathematical derivative of the charge Q versus voltage was well in line with the observed Cm changes. Based on these parameters a turnover rate of 500 molecules sucrose/s was calculated. In contrast to gating currents of voltage dependent-potassium channels the analysis of ZmSUT1-derived presteady-state currents in the absence of sucrose (I = Q/τ) was sufficient to predict ZmSUT1 transport-associated currents.
Conclusions: Taken together our results indicate that in the absence of sucrose, ‘trapped’ protons move back and forth between an outer and an inner site within the transmembrane domains of ZmSUT1. This movement of protons in the electric field of the membrane gives rise to the presteady-state currents and in turn to Cm changes. Upon application of external sucrose, protons can pass the membrane turning presteady-state into transport currents.
Das extrem thermophile Eubakterium Thermus thermophilus ist in den letzten Jahren zu einem Modell für thermophile Organismen geworden und verdankt seinen Statuszum Teil seiner hohe Wachstumsrate, den guten Zellerträgen und der konstitutivenExpression eines natürlichen Kompetenzapparates, der seine genetische Manipulation ermöglicht. Die Verfügbarkeit von kompatiblen Plasmiden und bis zu vier thermostabilen Antibiotikaresistenzmarkern konnten den Wert des Organismus in Hinblick auf biotechnologische Anwendungen noch weiter steigern und tatsächlich besteht nach wie vor ein ungebrochenes Interesse an der Struktur- und Funktionsaufklärung thermophiler Proteine. Der Focus der hier vorliegenden Arbeit richtete sich auf eine der insgesamt zwei terminalen Oxidasen der Atmungskette von T. thermophilus, die Cytochrom ba3 Oxidase. Es wurden verschiedene rekombinante Varianten des Proteins, die sich hinsichtlich der Position und Länge des verwendeten Histidin-Tags unterschieden, kloniert, exprimiert und aufgereinigt. Das Einfügen eines internen His12-Tags in einen periplasmatischen Loop zwischen den Transmembranhelices IV und V führte zu einer rekombinanten Version der Oxidase, die in ihren Eigenschaften dem nativen Wildtyp entsprach und sich durch die in dieser Arbeit etablierte Aufreinigungsstrategie relativ schnell, in guten Ausbeuten und hoher Reinheit aufreinigen ließ. Weiterhin konnten verschiedene Punktmutationen von möglicherweise am Elektronentransfer beteiligten Aminosäureresten generiert und die resultierenden Proteine aufgereinigt und über ihre enzymatische Aktivität charakterisiert werden. Eine weiterführende Charakterisierung der Mutanten erfolgte im Rahmen einer Kooperation und ist bisher noch nicht abgeschlossen. Das Herzstück dieser Arbeit machte jedoch die Definition der Transkriptionseinheit der Cytochrom ba3 Oxidase und die sich daraus ergebenden Fragestellungen aus. So konnte gezeigt werden, dass das ba3 Operon zusätzlich zu den Strukturgenen der dort kodierten Untereinheiten noch mindestens ein, höchst wahrscheinlich jedoch zwei zusätzliche Gene enthält: cbaX und cbaY. Bioinformatische Charakterisierungen ordneten CbaY der diversen Gruppe von sekundären Transportern zu, und es konnte experimentell gezeigt werden, dass seine Anwesenheit für die Expression der Cytochrom ba3 Oxidase förderlich ist. CbaX hingegen konnte über die durchgeführte Homologiesuche keine Funktion zugeordnet werden; uncharakterisierte Homologe waren einzig in der Thermaceae Gruppe zu finden. Durch Deletions- und Komplementationsstudien konnte dem Protein eine entscheidende Rolle in der Assemblierung der ba3 Oxidase bescheinigt werden. CbaX scheint eine zentrale Aufgabe bei der Häm a Insertion in Untereinheit I zu spielen und könnte, ohne Sequenzhomologie aufzuweisen, die Rolle des Surf1-Proteins übernehmen, welches in den sequenzierten Organismen der Thermaceae Gruppe nicht konserviert ist. Die homologe Expression und Aufreinigung von CbaX führte nicht zur erwarteten Ausbeute und Reinheit des Proteins, konnte aber durch immunologische Experimente eine potentielle Interaktion von CbaX und der Cytochrom ba3 Oxidase nachweisen.
Background: ClC-7 is a ubiquitous transporter which is broadly expressed in mammalian tissues. It is implied in the pathogenesis of lysosomal storage disease and osteopetrosis. Because of its endosomal/lysosomal localization it is still poorly characterized. Methodology/Principal Findings: An electrophysiological characterization of rat ClC-7 using solid-supported membrane-based electrophysiology is presented. The measured currents show the characteristics of ClC-7 and confirm its function as a Cl−/H+-antiporter. We have used rat ClC-7 in CHO cells as a model system to investigate the functionality and cellular localization of the wt transporter and its variant G213R ClC-7 which is the analogue of human G215R ClC-7 responsible for autosomal dominant osteopetrosis type II. Our study shows that rat G213R ClC-7 is functional but has a localization defect in CHO cells which prevents it from being correctly targeted to the lysosomal membrane. The electrophysiological assay is tested as a tool for drug discovery. The assay is validated with a number of drug candidates. It is shown that ClC-7 is inhibited by DIDS, NPPB and NS5818 at micromolar concentrations. Conclusions/Significance: It is suggested that the scenario found in the CHO model system also applies to the human transporter and that mislocalization rather than impaired functionality of G215R ClC-7 is the primary cause of the related autosomal dominant osteopetrosis type II. Furthermore, the robust solid-supported membrane-based electrophysiological assay is proposed for rapid screening for potential ClC-7 inhibitors which are discussed for treatment of osteoporosis.
The continuous progress in the structural and functional characterization of aquaporins increasingly attracts attention to study their roles in certain mammalian diseases. Although several structures of aquaporins have already been solved by crystallization, the challenge of producing sufficient amounts of functional proteins still remains. CF (cell free) expression has emerged in recent times as a promising alternative option in order to synthesize large quantities of membrane proteins, and the focus of this report was to evaluate the potential of this technique for the production of eukaryotic aquaporins. We have selected the mouse aquaporin 4 as a representative of mammalian aquaporins. The protein was synthesized in an E. coli extract based cell-free system with two different expression modes, and the efficiencies of two modes were compared. In both, the P-CF (cell-free membrane protein expression as precipitate) mode generating initial aquaporin precipitates as well as in the D-CF (cell-free membrane protein expression in presence of detergent) mode, generating directly detergent solubilized samples, we were able to obtain mg amounts of protein per ml of cell-free reaction. Purified aquaporin samples solubilized in different detergents were reconstituted into liposomes, and analyzed for the water channel activity. The calculated Pf value of proteoliposome samples isolated from the D-CF mode was 133 µm/s at 10°C, which was 5 times higher as that of the control. A reversible inhibitory effect of mercury chloride was observed, which is consistent with previous observations of in vitro reconstituted aquaporin 4. In this study, a fast and convenient protocol was established for functional expression of aquaporins, which could serve as basis for further applications such as water filtration.
