Karl Nauwerck in Sizilien : eine Edition zweier früher sozialkritischer Bildungsreiseberichte
(2009)
The representation of music in literature is often conceived in terms of a 'paragone', a debate over which of the arts ranks highest. In this context, literature is commonly spoken of as imitative or emulative of the music it is trying to represent. This premise informs even the most recent terminologies of literary scholars describing the intermedial relationship of music and literature systematically. Using the narratological concepts of 'showing vs. telling' to describe musical representation in literature, as proposed by the "Handbuch Literatur & Musik", this article argues that such a view is based on a misleading assumption about literary imitation and leads to constrained possibilities of hermeneutical interpretation. After a systematic reconstruction of the proposed terminology and a proposal for its modification, an exemplary analysis of Helmut Krausser's novel "Melodien" ["Melodies", 1993] will serve to demonstrate how a refined conception of the terminology can help to bring about more precise interpretations.

Biophysical investigation of the ligand-induced assembling of the human type I interferon receptor
(2005)
Type I interferons (IFNs) elicit antiviral, antiproliferative and immunmodulatory responses through binding to a shared receptor consisting of the transmembrane proteins ifnar1 and ifnar2. Differential signaling by different interferons – in particular IFNalpha´s and IFNbeta – suggest different modes of receptor engagement. In this work either single ligand-receptor interactions or the formation of the extracellular part of a signaling complex were investigated referring to thermodynamics, kinetics, stoichiometry and structural organization. Initially an expression and purification strategy for the extracellular domain of ifnar1 (ifnar1-EC) using Sf9 insect cells yielding in mg amounts of glycosylated protein was established. Using reflectometric interference spectroscopy (RIfS) the interactions between IFNalpha2/beta and ifnar1-EC and ifnar2-EC was studied in order to understand the individual energetic contributions within the ternary complex. For IFNalpha2 a Kd of 5 µM for the interaction with ifnar1-EC was determined. Substantially tighter binding of IFNbeta with both ifnar2-EC and ifnar1-EC compared to IFNalpha2 was observed. For neither IFNalpha2 nor IFNbeta stabilization of the complex with ifnar1-EC in presence of soluble ifnar2-EC was detectable. In addition, no direct interaction between ifnar2 and ifnar1 was could be shown. Thus, stem-stem interactions between the extracellular domains of ifnar1 and ifnar2 do not seem to play a role for ternary complex formation. Furthermore, ligand-induced cross-talk between ifnar1-EC and ifnar2-EC being tethered onto solid-supported, fluid lipid bilayers was investigated by RIfS and total internal reflection fluorescence spectroscopy. A very stable binding of IFNalpha2 at high receptor surface concentrations was observed with an apparent kd approximately 200-times lower than for ifnar2-EC alone. This apparent kd was strongly dependent on the surface concentration of the receptor components, suggesting kinetic rather than static stabilization, which was corroborated by competition experiments. These results indicate that signaling is activated by transient cross-talk between ifnar1 and ifnar2, which is by several orders of magnitude more efficiently engaged by IFNbeta than by IFNalpha2. With respect to differential recognition of different IFNs ifnar1-EC was dissected into sub-fragments containing different of the four Ig-like domains. The appropriate folding and glycosylation of these proteins, also purified in mg amounts were confirmed by SDS-PAGE, size exclusion chromatography and CD-spectroscopy. Surprisingly, only one construct containing all three N-terminal Ig-like domains was active in terms of ligand binding, indicating that these domains were required. Competitive binding of IFNalpha2 and IFNbeta to both this fragment and ifnar1-EC was demonstrated. Cellular binding assays with different fragments, however, highlight the key role of the membrane-proximal Ig-like domain for the formation of an in situ IFN-receptor complex and the ensuing signal activation. Even substitution with Ig-like domains from homologous cytokine receptors did not restore high-affinity ligand binding. Receptor assembling analysis on supported lipid bilayer revealed that appropriate orientation of the receptor is required, which is controlled by the membrane-proximal Ig-domain. All results indicate that differential signalling is encoded by the efficiency of signalling complex formation, which is controlled by the binding affinity of IFNs to the extracellular domains of ifnar1 and 2.
Microbial rhodopsins are omnipresent on Earth, however the vast majority of them remain uncharacterized. Here we describe a new rhodopsin group from cold-adapted organisms and cold environments, such as glaciers, denoted as CryoRhodopsins (CryoRs). Our data suggest that CryoRs have dual functionality switching between inward transmembrane proton translocation and photosensory activity, both of which can be modulated with UV light. CryoR1 exhibits two subpopulations in the ground state, which upon light activation lead to transient photocurrents of opposing polarities. A distinguishing feature of the group is the presence of a buried arginine residue close to the cytoplasmic face of its members. Combining single-particle cryo-electron microscopy and X-ray crystallography with the rhodopsin activation by lit, we demonstrate that the arginine stabilizes a UV-absorbing intermediate of an extremely slow CryoRhodopsin photocycle. Together with extensive spectroscopic characterization, our investigations on CryoR1 and CryoR2 proteins reveal mechanisms of photoswitching in the newly identified group and demonstrate principles of the adaptation of these rhodopsins to low temperatures.Microbial rhodopsins are omnipresent on Earth, however the vast majority of them remain uncharacterized.
Cryptocurrencies have received growing attention from individuals, the media, and regulators. However, little is known about the investors whom these financial instruments attract. Using administrative data, we describe the investment behavior of individuals who invest in cryptocurrencies with structured retail products. We find that cryptocurrency investors are active traders, prone to investment biases, and hold risky portfolios. In line with attention effects and anticipatory utility, we find that the average cryptocurrency investor substantially increases log-in and trading activity after his or her first cryptocurrency purchase. Our results document which investors are more likely to adopt new financial products and help inform regulators about investors' vulnerability to cryptocurrency investments.

Transposable elements (TEs) are replicating genetic elementst hat comprise up to 50% of mammalian genomes. A specific class of TEs are retrotransposons that proliferate by transcription into a RNA intermediate, followed by genomic reintegration into another locus (so called “copy & paste” mechanism). Due to the lack of removal mechanisms and very rare parallel insertions, the presence of TE insertions at ortholgous genomic loci in multiple taxa provides a virtually homoplasy free phylogenetic marker. So far, developing phylogenetically informative markers from TE insertions has been a tedious work of testing hundreds of putative candidate loci in a trial-and error approach with low success rate. Hence, phylogenetic studies using TE insertions were often limited to a few dozen markers.
Recently, genome sequencing of multiple species using reference-mapping allowed the identification of genome-scale datasets of TE insertions. and made the ad-hoc development of phylogenetic informative markers possible. However, genome scale TE detection methods have rarely been applied to non model organisms in which data availability and quality is comparably limited. In this thesis, I developed the TeddyPi pipeline (TE detection and discovery for phylogenetic inference), a software tool that made it possible to obtain reliable genome-scale TE insertion data from low-coverage genomes. This was achieved by integrating the data from multiple TE and structural variation callers as well as applying a stringent filtering pipeline to exclude low-quality insertion calls. Whole-genome sequencing datasets of bears (Ursidae) and baleen whales (Mysticeti) were used to apply TE based phylogenetic inference and evaluate the method in comparison to sequence-based phylogenomic analyses.
In the bear genomes, TeddyPi identified 150,513 high-quality transposable element (TE) insertions, which allowed me to reconstruct the evolutionary history of bears despite extensive phylogenetic conflict (Lammers et al., 2017). The large number of detected TE insertions made also detailed network analyses possible that visualize the phylogenetic conflict. Experimental polymerase chain reaction (PCR) assays validated up to 93 % of the computationally identified TE loci and demonstrated the high accuracy of the dataset underlying the phylogenetic analyses.
Second, I present the initial genome sequencing of six baleen whales and a detailed investigation of their evolutionary history using TE insertions and established sequence-based phylogenomic methods. The taxon sampling of baleen whales included iconic species like the blue whale (Balaneoptera musculus) or the humpback whale (Megaptera novaengliae) (Árnason et al., 2018). A sequence-based reconstruction of the baleen whale species tree solved the long-debated phylogenetic position of the gray whale (Echrichtius robustus) within rorquals (Balaneopteridae) for the first time with high statistical support. Furthermore, the genome data made it possible to identify large extent of phylogenetic conflict for divergences during the radiation of rorquals that occurred 7-10 million years ago (Ma).
The phylogenomic analyses of 91,589 TE insertions in the whale genomes confirmed the sequence-based topology (Lammers et al., 2019). The quantification of phylogenetic signals obtained from the TE insertions revealed a high degree of discordance for the divergence of the gray whale and rorquals. Despite the large genome-scale dataset, statistical tests showed only marginal support for a bifurcating divergence of gray whales and the rorqual species. The limited statistical support for a strictly bifurcating tree obtained from genome-scale datasets of thousands of markers demonstrates the importance for including phylogenetic networks for displaying evolutionary divergences.
In conclusion, this thesis shows that identification of TE insertions from whole-genome resequencing provides plentiful and accurate phylogenomic markers. For the application in non model organisms, I provide a easy-to-use software to integrate multiple datasets from TE and structural variation callers in order to obtain reliable and ascertainment-bias free datasets. Detecting genome-scale datasets of TE insertions in two case studies demonstrates the applicability of this marker system for phylogenetic reconstruction and inferring phylogenetic conflict.
Retrophylogenomics in rorquals indicate large ancestral population sizes and a rapid radiation
(2019)
Background: Baleen whales (Mysticeti) are the largest animals on earth and their evolutionary history has been studied in detail, but some relationships still remain contentious. In particular, reconstructing the phylogenetic position of the gray whales (Eschrichtiidae) has been complicated by evolutionary processes such as gene flow and incomplete lineage sorting (ILS). Here, whole-genome sequencing data of the extant baleen whale radiation allowed us to identify transposable element (TE) insertions in order to perform phylogenomic analyses and measure germline insertion rates of TEs. Baleen whales exhibit the slowest nucleotide substitution rate among mammals, hence we additionally examined the evolutionary insertion rates of TE insertions across the genomes.
Results: In eleven whole-genome sequences representing the extant radiation of baleen whales, we identified 91,859 CHR-SINE insertions that were used to reconstruct the phylogeny with different approaches as well as perform evolutionary network analyses and a quantification of conflicting phylogenetic signals. Our results indicate that the radiation of rorquals and gray whales might not be bifurcating. The morphologically derived gray whales are placed inside the rorqual group, as the sister-species to humpback and fin whales. Detailed investigation of TE insertion rates confirm that a mutational slow down in the whale lineage is present but less pronounced for TEs than for nucleotide substitutions.
Conclusions: Whole genome sequencing based detection of TE insertions showed that the speciation processes in baleen whales represent a rapid radiation. Large genome-scale TE data sets in addition allow to understand retrotransposition rates in non-model organisms and show the potential for TE calling methods to study the evolutionary history of species.
Compared to sequence analyses, phylogenetic reconstruction from transposable elements (TEs) offers an additional perspective to study evolutionary processes. However, detecting phylogenetically informative TE insertions requires tedious experimental work, limiting the power of phylogenetic inference. Here, we analyzed the genomes of seven bear species using high throughput sequencing data to detect thousands of TE insertions. The newly developed pipeline for TE detection called TeddyPi (TE detection and discovery for Phylogenetic Inference) obtained 150,513 high-quality TE insertions in the genomes of ursine and tremarctine bears. By integrating different TE insertion callers and using a stringent filtering approach, the TeddyPi pipeline produced highly reliable TE insertion calls, which were confirmed by extensive in vitro validation experiments. Screening for single nucleotide substitutions in the flanking regions of the TEs show that these substitutions correlate with the phylogenetic signal from the TE insertions. Our phylogenomic analyses show that TEs are a major driver of genomic variation in bears and enabled phylogenetic reconstruction of a well-resolved species tree, even with strong signals for incomplete lineage sorting and introgression. The analyses show that the Asiatic black, sun and sloth bear form a monophyletic clade. TeddyPi is open source and can be adapted to various TE and structural variation callers. The pipeline makes it easy to confidently extract thousands of TE insertions even from low coverage genomes of non-model organisms, opening new possibilities for biologists to study phylogenies, evolutionary processes as well as rates and patterns of (retro-)transposition and structural variation.

Phylogenetic analyses of nuclear and mitochondrial genomes have shown that polar bears captured the mitochondrial genome of brown bears some 160,00 years ago. This hybridization event likely led to an extinction of the original polar bear mitochondrial genome. However, parts of the mitochondrial DNA occasionally integrates into the nuclear genome, forming pseudogenes called numts (nuclear mitochondrial integrations). Screening the polar bear genome for numts, we identified only 13 such integrations. Analyses of whole-genome sequences from additional polar bears, brown and American black bears as well as the giant panda indicates that the discovered numts entered the bear lineage before the initial ursid radiation some 14 million years ago. Our findings suggests a low integration rate of numts in the bear lineage and a complete loss of the original polar bear mitochondrial genome.

Our findings suggest low integration rates of numts in bears and a loss of the original polar bear mitochondrial genome.

TO CONSIDER THE IMPACT OF UNCERTAIN MARKET ENVIRONMENTS ON INAND OUTSOURCING DECISIONS, WE INTRODUCE A REAL OPTIONS BASED DECISION SUPPORT MODEL. WE APPLY THE MODEL TO AN IT INFRASTRUCTURE OUTSOURCING DECISION AND DETERMINE - BASED ON COST SAVINGS RESULTING FROM OUTSOURCING AND OPTION VALUES ACCOUNTING FOR UNCERTAINTY - DIFFERENT “TRIGGER” OUTPUT VOLUMES WHICH INDICATE IF IN- OR OUTSOURCING IS PREFERABLE. FINALLY WE SHOW THAT THE MODEL CAN ALSO BE TRANSFERRED TO SOURCING DECISIONS OF TRANSACTION BASED BUSINESS PROCESSES LIKE CLEARING AND SETTLEMENT OF SECURITIES.
The privatization of Old Age and Survivors Insurance and Disability Insurance was a top priority on president Bush’s domestic political agenda. Although Bush’s reform initiative has failed and president Obama has declared not to privatize social security, the system of public old age security in the United States is still in crisis, mainly because of demographic factors and the ensuing financial problems but also because of the recent and deep economic recession in the United States. This article reviews the initiative of the Bush-Administration to partially privatize social security and analyzes the main objectives behind Bush policy as well as the main arguments against and obstacles to it. By placing Bush politics of privatizing social security in a broader context of comparative welfare state reform, this article discusses the consequences of privatizing social security systems on equality and poverty, as well as on the legitimacy of the political system in general.
Since the early 1960s ethno-territorial movements have increasingly challenged established patterns of political integration in western democracies. The purpose of this paper is to compare the formation and different paths of development of such nationalist movements in Quebec (Canada) and Corsica (France). The paper presents a brief outline of an argument more fully developed in a study on the two cases. After a short introduction I will first discuss theoretical aspects of minority nationalism. Secondly, I will try to give an overview over major findings and conclusions of my study.
The main purpose of the paper is to explain the divergent paths of development of ethno-territorial protest movements in modern democratic political systems. By focusing on the interaction between these movements and the state, the different systems of accommodation between the relevant regional and central elites will be analyzed. The study concentrates on the case studies of Québec (Canada) and Corsica (France). The paper is divided into three parts. The first part describes the traditional systems of accommodation in France and Canada. The second part is focused on the process of socio-economic modernization in the 1950s and 1960s in those countries that threatened the established patterns of elite accommodation. The third part deals with the consequences for the established patterns of elite-accommodation and new concepts of territorial management that the central states tried to establish. By looking at the different degrees of centralization and decentralization in the mentioned political systems, the question of access to the political system by new social and political actors will be discussed in detail.
Taxation and tax policy reform appears on the political agenda in most advanced welfare states in Europe and North America. Of course studies of taxation and tax policy are nothing new and have existed ever since people have paid taxes. The current work is situated in the context of the future of the welfare state and the reinforced international economic and political integration referred to as "globalization." The purpose of this paper is to analyze how globalization is affecting tax policy in advanced welfare states. In comparing the evolution of tax policy in Canada with those in the United States, Germany and Sweden from 1960 to 1995, I will try to review the conventional antiglobalization thesis, i.e., that globalization leads to a "race to the bottom" in revenue and expenditures policies, or as others have called it, a "beggar the neighbour policy" (Tanzi and Bovenberg 1990, 187). ... Conclusion: The empirical data and theoretical models clearly show that globalization is one relatively minor factor among many that explain tax policy reforms. And even that limited influence is mediated by domestic political systems, institutions and constellations of actors. As the data has shown, the conventional globalization thesis of a race to the bottom is not borne out. Tax rates and tax revenues are still increasing, despite the ongoing trend toward international trade integration. Countervailing pressures like the high cost of welfare programs, different parties in government, strong labour unions, and institutional veto players counteract the pressure of globalization on tax policy. As for the future of taxation in Canada, it is more likely to be one of gradual evolution than radical change. Although the data don’t show any downward pressure on tax rates and tax revenues comparatively speaking, there are at least four key factors in Canada that are likely to put pressure on future tax rates, although regional political dynamics and the workings of fiscal federalism suggest that tax reductions will be a higher priority in some provinces than others (Hale 2002). First, neoliberalism will continue to shape fiscal and tax policy, including the role of the tax system in delivering social policies and programs in most parts of Canada. Second, governments that seek to define their own economic and social priorities rather than simply react to events beyond their borders will have to exercise centralized control over budgetary policies and spending levels if they hope to foster the economic growth needed to finance social services in the context of Canada’s changing demographics. Third, the ability of governments to combine the promotion of economic growth and higher living standards will be closely linked to their ability to develop a workable division of responsibilities among federal and provincial governments and with other national governments. Finally, the diffusion of new technologies will continue to transform national and regional economies while giving individuals greater opportunity to avoid government and tax regulations that run contrary to their perceived interests and values. This discussion of determinants that shape tax policy reform has shown that successful management of fiscal and tax policy requires a capacity to set priorities; adapt to changing circumstances; and build a consensus that enables competing economic, social, regional and ideological interests to identify their own well-being in the broader political and economic environment. Tax policy is shaped by many political, economic and social determinants. As Geoffrey Hale correctly concludes, "it should not be surprising if the tax system stubbornly refuses to confirm either economic theories or political ideologies, but reflects past decisions and the policy tradeoffs of the political process" (2002, 71). The notion of tax policy being driven by globalization and forces associated with globalization (both positive and negative) is simply not borne by the facts.
This thesis primarily investigates an (hitherto unnoticed) agreement alternation between Romance and Germanic in D>N>&>N constructions (e.g. “these walls and ceiling”). While Romance exhibits left conjunct agreement, Germanic shows morphologically resolved agreement on the determiner, i.e. the phi-feature mismatching conjuncts can only be licensed if a syncretic form is available. To handle these data the author suggests a theory in which coordination is syntactically and morphologically unspecified and multiple agree is a general option. Infelicitous derivations are ruled out by interface conditions on the semantic and the phonological well formedness. The complete results of the corpus research conducted to deliver a sound empirical basis for the phenomena investigated in this thesis can be found in the appendix.
This detailed, meticulous ethnographic study on mobile phone use among Nuba students at the University of Khartoum in Sudan, distinguishes itself from other studies by taking a focused look at the linguistic content of mobile phone interactions via text-messaging, portraying it as a site for the expression of personalized and affective language. While men and women appear to be equally aggressive consumers and producers of text-message poetry, women are formally discouraged in using the phone for relations that go beyond the publicly acceptable norms of 'keeping in touch' and making arrangements. Nonetheless, women use it for such purposes and many manage it discreetly, showing how this technology can serve to subvert discursive norms on gender and marriage. The mobile phone in Sudan enhances individual autonomy over interactions, making possible the extension and creation of social spaces. It simultaneously enlarges private space and trespasses into public space. Poetic themes and language, previously limited to elite producers - those both more literate and who had control over mass media domains, radio and newspapers - are exposed to anonymous recipients, who draw from, copy or forward them in continuous circulation, thereby staking a claim in the public sphere. Similarly, the mobile phone serves as a site for the exercise of several layers of identity in negotiation, and reflects or creates alternative identities and the contestation of existing discourses, communities in physical space and notions of belonging.
Background: Glaucoma is a neurodegenerative disease, leading to thinning of the retinal nerve fibre layer (RNFL). The exact influence of ocular, cardiovascular, morphometric, lifestyle and cognitive factors on RNFL thickness (RNFLT) is unknown and was analysed in a subgroup of the Gutenberg Health Study (GHS).
Methods: Global peripapillary RNFLT was measured in 3224 eyes of 1973 subjects (49% female) using spectral-domain optical coherence tomography (SD-OCT). The association of age, sex, ocular, cardiovascular, morphometric, lifestyle and cognitive factors on RNFLT was analysed using Pearson correlation coefficient and fitting a linear mixed model.
Results: In the univariable analysis highest correlations were found for axial length (r = -0.27), spherical equivalent (r = 0.24), and glaucoma (r = -0.15) (p<0.0001, respectively). Other significant correlations with RNFLT were found for age, sex, intraocular pressure, systemic hypertension and systolic blood pressure, previous eye surgery, cholesterol, homocysteine, history of coronary artery disease, history of myocardial infarction, apnoea, diabetes and alcohol intake, p<0.05, respectively. Body length, body weight, BMI, diastolic blood pressure, blood glucose, HbA1c, history of apoplexy, cognitive function, peripheral artery disease, tinnitus, migraine, nicotine intake, central corneal thickness, and pseudophakia were not significantly correlated with RNFLT. The regression model revealed a significant relationship between RNFLT and age in decades (p<0.02), spherical equivalent (p<0.0001), axial length (p<0.0001), glaucoma (p<0.0001), tinnitus (p = 0.04), apnoea (p = 0.047), homocysteine (p = 0.05) and alcohol intake >10g/d for women and >20g/d for men (p = 0.02). Glaucoma, apnoea, higher homocysteine, higher alcohol intake and higher axial length as well as age were related to decreased RNFLT while higher spherical equivalent or history for tinnitus were related to thicker RNFL.
Conclusion: RNFLT is related to age, ocular parameters and lifestyle factors. Considering these parameters in normative databases could improve the evaluation of peripapillary RNFLT. It is necessary to evaluate if a reduction of alcohol intake as well as the therapy of apnea or high homocysteine levels could positively influence RNFLT.
Importance: The entry of artificial intelligence into medicine is pending. Several methods have been used for the predictions of structured neuroimaging data, yet nobody compared them in this context.
Objective: Multi-class prediction is key for building computational aid systems for differential diagnosis. We compared support vector machine, random forest, gradient boosting, and deep feed-forward neural networks for the classification of different neurodegenerative syndromes based on structural magnetic resonance imaging.
Design, setting, and participants: Atlas-based volumetry was performed on multi-centric T1-weighted MRI data from 940 subjects, i.e., 124 healthy controls and 816 patients with ten different neurodegenerative diseases, leading to a multi-diagnostic multi-class classification task with eleven different classes.
Interventions: N.A.
Main outcomes and measures: Cohen’s kappa, accuracy, and F1-score to assess model performance.
Results: Overall, the neural network produced both the best performance measures and the most robust results. The smaller classes however were better classified by either the ensemble learning methods or the support vector machine, while performance measures for small classes were comparatively low, as expected. Diseases with regionally specific and pronounced atrophy patterns were generally better classified than diseases with widespread and rather weak atrophy.
Conclusions and relevance: Our study furthermore underlines the necessity of larger data sets but also calls for a careful consideration of different machine learning methods that can handle the type of data and the classification task best.
