We present measurements of the azimuthal dependence of charged jet production in central and semi-central sNN−−−√ = 2.76 TeV Pb-Pb collisions with respect to the second harmonic event plane, quantified as vch jet2. Jet finding is performed employing the anti-kT algorithm with a resolution parameter R = 0.2 using charged tracks from the ALICE tracking system. The contribution of the azimuthal anisotropy of the underlying event is taken into account event-by-event. The remaining (statistical) region-to-region fluctuations are removed on an ensemble basis by unfolding the jet spectra for different event plane orientations independently. Significant non-zero vch jet2 is observed in semi-central collisions (30-50\% centrality) for 20 < pch jetT < 90 GeV/c. The azimuthal dependence of the charged jet production is similar to the dependence observed for jets comprising both charged and neutral fragments, and compatible with measurements of the v2 of single charged particles at high pT. Good agreement between the data and predictions from JEWEL, an event generator simulating parton shower evolution in the presence of a dense QCD medium, is found in semi-central collisions.
The production of prompt charmed mesons D0, D+ and D∗+, and their antiparticles, was measured with the ALICE detector in Pb-Pb collisions at the centre-of-mass energy per nucleon pair, sNN−−−√, of 2.76 TeV. The production yields for rapidity |y|<0.5 are presented as a function of transverse momentum, pT, in the interval 1-36 GeV/c for the centrality class 0-10% and in the interval 1-16 GeV/c for the centrality class 30-50%. The nuclear modification factor RAA was computed using a proton-proton reference at s√=2.76 TeV, based on measurements at s√=7 TeV and on theoretical calculations. A maximum suppression by a factor of 5-6 with respect to binary-scaled pp yields is observed for the most central collisions at pT of about 10 GeV/c. A suppression by a factor of about 2-3 persists at the highest pT covered by the measurements. At low pT (1-3 GeV/c), the RAA has large uncertainties that span the range 0.35 (factor of about 3 suppression) to 1 (no suppression). In all pT intervals, the RAA is larger in the 30-50% centrality class compared to central collisions. The D-meson RAA is also compared with that of charged pions and, at large pT, charged hadrons, and with model calculations.
The centrality dependence of the charged-particle pseudorapidity density measured with ALICE in Pb-Pb collisions at sNN−−−√ over a broad pseudorapidity range is presented. This Letter extends the previous results reported by ALICE to more peripheral collisions. No strong change of the charged-particle pseudorapidity density distributions with centrality is observed, and when normalised to the number of participating nucleons in the collisions, the evolution over pseudorapidity with centrality is likewise small. The broad pseudorapidity range allows precise estimates of the total number of produced charged particles which we find to range from 162±22 (syst.) to 17170±770 (syst.) in 80-90% and 0-5 central collisions, respectively. The total charged-particle multiplicity is seen to approximately scale with the number of participating nucleons in the collision. This suggests that hard contributions to the charged-particle multiplicity are limited. The results are compared to models which describe dNch/dη at mid-rapidity in the most central Pb-Pb collisions and it is found that these models do not capture all features of the distributions.
Visual processing begins at the first synapse of the visual system. In the mouse retina, three different types of photoreceptors provide input to 14 bipolar cell (BC) types. Classically, most BC types are thought to contact all cones within their dendritic field; ON BCs would contact cones exclusively via so-called invaginating synapses, while OFF BCs would form basal synapses. By mining publically available electron microscopy data, we discovered interesting violations of these rules of outer retinal connectivity: ON BC type X contacted only ~20% of the cones in its dendritic field and made mostly atypical non-invaginating contacts. Types 5T, 5O and 8 also contacted fewer cones than expected. In addition, we found that rod BCs received input from cones, providing anatomical evidence that rod and cone pathways are interconnected in both directions. This suggests that the organization of the outer plexiform layer is more complex than classically thought.
Wie Rolf Parr in seinem Aufsatz 'Liminale und andere Übergänge. Theoretische Modellierung von Grenzzonen, Normalitätsaspekten, Schwellen, Übergängen und Zwischenräumen in Literatur- und Kulturwissenschaft' deutlich macht, ist die Intertextualitäts- und Intermedialitätstheorie, die er im Anschluss an die Arbeiten Michel Foucaults und Jürgen Links vertritt, wesentlich von einem Moment der Grenzüberschreitung bestimmt. An die Stelle klar konturierter Grenzen treten Schwellen als "räumlichtopographische Zonen der Unentschiedenheit", die zugleich als zeitliche Erinnerungsschwellen fungieren. Parr richtet im Rekurs auf Foucault den Blick nicht allein auf diskursive Grenzen der Sagbarkeit durch Ausschlussmechanismen, Verbote etc. Er macht zugleich auf Foucaults frühes Konzept der Heterotopie aufmerksam, wo dieser Grenzziehungen auf bestimmte Raumstrukturen bezieht. Parrs eigenes Interesse liegt in diesem Zusammenhang in der Überführung der diskurstheoretischen Arbeiten Foucaults in eine Interdiskurstheorie, die eben die Schwellen einzelner Diskurse zu überschreiten hätte. Ich möchte hier einen anderen Akzent setzen und die Bedeutung von Schwellenerfahrungen bei Foucault selbst herausarbeiten. Ich konzentriere mich dabei zunächst auf den Begriff des historischen Aprioris aus 'Die Ordnung der Dinge', um daran anschließend auf den Begriff der Heterotopie einzugehen, der die Entstehung der 'Ordnung der Dinge' in den sechziger Jahren in gewisser Weise begleitet und komplementiert. Der Vergleich von Foucaults Schwellendenken mit dem Walter Benjamins soll zugleich erlauben, das Thema des Liminalen im Sinne Parrs als ein Grundmotiv von Foucaults Denken auszumachen.
Bipolar disorder (BD) is a genetically complex mental illness characterized by severe oscillations of mood and behavior. Genome-wide association studies (GWAS) have identified several risk loci that together account for a small portion of the heritability. To identify additional risk loci, we performed a two-stage meta-analysis of >9 million genetic variants in 9,784 bipolar disorder patients and 30,471 controls, the largest GWAS of BD to date. In this study, to increase power we used ~2,000 lithium-treated cases with a long-term diagnosis of BD from the Consortium on Lithium Genetics, excess controls, and analytic methods optimized for markers on the Xchromosome. In addition to four known loci, results revealed genome-wide significant associations at two novel loci: an intergenic region on 9p21.3 (rs12553324, p = 5.87×10-9; odds ratio = 1.12) and markers within ERBB2 (rs2517959, p = 4.53×10-9; odds ratio = 1.13). No significant X-chromosome associations were detected and X-linked markers explained very little BD heritability. The results add to a growing list of common autosomal variants involved in BD and illustrate the power of comparing well-characterized cases to an excess of controls in GWAS.
Bears are iconic mammals with a complex evolutionary history. Natural bear hybrids and studies of few nuclear genes indicate that gene flow among bears may be more common than expected and not limited to the closely related polar and brown bears. Here we present a genome analysis of the bear family with representatives of all living species. Phylogenomic analyses of 869 mega base pairs divided into 18,621 genome fragments yielded a well-resolved coalescent species tree despite signals for extensive gene flow across species. However, genome analyses using three different statistical methods show that gene flow is not limited to closely related species pairs. Strong ancestral gene flow between the Asiatic black bear and the ancestor to polar, brown and American black bear explains numerous uncertainties in reconstructing the bear phylogeny. Gene flow across the bear clade may be mediated by intermediate species such as the geographically wide-spread brown bears leading to massive amounts of phylogenetic conflict. Genome-scale analyses lead to a more complete understanding of complex evolutionary processes. The increasing evidence for extensive inter-specific gene flow, found also in other animal species, necessitates shifting the attention from speciation processes achieving genome-wide reproductive isolation to the selective processes that maintain species divergence in the face of gene flow.
Phylogenetic analyses of nuclear and mitochondrial genomes have shown that polar bears captured the mitochondrial genome of brown bears some 160,00 years ago. This hybridization event likely led to an extinction of the original polar bear mitochondrial genome. However, parts of the mitochondrial DNA occasionally integrates into the nuclear genome, forming pseudogenes called numts (nuclear mitochondrial integrations). Screening the polar bear genome for numts, we identified only 13 such integrations. Analyses of whole-genome sequences from additional polar bears, brown and American black bears as well as the giant panda indicates that the discovered numts entered the bear lineage before the initial ursid radiation some 14 million years ago. Our findings suggests a low integration rate of numts in the bear lineage and a complete loss of the original polar bear mitochondrial genome.
The interaction between the Heat Shock Proteins 70 and 40 is at the core of the ATPase regulation of the chaperone machinery that maintains protein homeostasis. However, the structural details of this fundamental interaction are still elusive and contrasting models have been proposed for the transient Hsp70/Hsp40 complexes. Here we combine molecular simulations based on both coarsegrained and atomistic models with co-evolutionary sequence analysis to shed light on this problem by focusing on the bacterial DnaK/DnaJ system. The integration of these complementary approaches resulted into a novel structural model that rationalizes previous experimental observations. We identify an evolutionary-conserved interaction surface formed by helix II of the DnaJ J-domain and a groove on lobe IIA of the DnaK nucleotide binding domain, involving the inter-domain linker.
Active transposable elements (TEs) may result in divergent genomic insertion and abundance patterns among conspecific populations. Upon secondary contact, such divergent genetic backgrounds can theoretically give rise to classical Dobzhansky-Muller incompatibilities (DMI), a way how TEs can contribute to the evolution of endogenous genetic barriers and eventually population divergence. We investigated whether differential TE activity created endogenous selection pressures among conspecific populations of the non-biting midge Chironomus riparius, focussing on a Chironomus-specific TE, the minisatellite-like Cla-element, whose activity is associated with speciation in the genus. Using an improved and annotated draft genome for a genomic study with five natural C. riparius populations, we found highly population-specific TE insertion patterns with many private insertions. A highly significant correlation of pairwise population FST from genome-wide SNPs with the FST estimated from TEs suggests drift as the major force driving TE population differentiation. However, the significantly higher Cla-element FST level due to a high proportion of differentially fixed Cla-element insertions indicates that segregating, i.e. heterozygous insertions are selected against. With reciprocal crossing experiments and fluorescent in-situ hybridisation of Cla-elements to polytene chromosomes, we documented phenotypic effects on female fertility and chromosomal mispairings that might be linked to DMI in hybrids. We propose that the inferred negative selection on heterozygous Cla-element insertions causes endogenous genetic barriers and therefore acts as DMI among C. riparius populations. The intrinsic genomic turnover exerted by TEs, thus, may have a direct impact on population divergence that is operationally different from drift and local adaptation.
