Introduction: The optimal treatment of patients with spinal infections remains a controversial topic. Within Europe, fundamentally different therapeutic concepts are found. Therefore, the aim of this study was to compare the outcome of patients who received surgical vs. antibiotic treatment alone for primary pyogenic spondylodiscitis in an international cohort analysis.
Materials and Methods: The retrospectively compiled databases of tertiary high-volume spine centers served as the baseline for this study. All documented cases of primary spondylodiscitis treated surgically and conservatively in the period of 2017-2022 were included and grouped according to the therapeutic concept: conservative vs. surgical treatment. Independent investigators collected the relevant clinical and radiological data. The primary endpoint of this study was mortality rate; secondary endpoints were relapse rate and persisting neurological deficit.
Results: A total of 392 patients were included in the analysis (155 females with a mean age of 68 years). Of these, 95 cases were treated conservatively (CoT) and 297 cases were treated surgically (SuT). There was no significant difference (p<0.01) related to patient’s disease characteristics: Lumbar was the main location (n=240, CoT 58/ SuT 182, p=0.97) followed by thoracic (n=70, CoT 24/ SuT 46, p=0,03) and cervical (n=47, CoT 7/ SuT 40, p=0.11) region. A multilocular spinal infection was present in 32 patients (CoT 3/ SuT 29, p=0.04). 181 cases (CoT 36/ SuT 145, p=0.06) presented with an epidural abscess. Neurological deficits were recorded in 100 cases (CoT 26/ SuT 74, p=0.63), and septic conditions in 88 cases (CoT 26/ SuT 62, p=0.19). Pre-existing conditions like Diabetes (p=0.57), renal failure (p= 0.97), hepatopathy (p= 0.15), malignoma (p=0.39) or i.v. drug abuse (p=0.93) did also not differ between the groups. The mortality rate of all conservatively treated was 24.2% (23 cases) and 6.7% (20 cases) in all surgically treated patients (p<0.001). A follow-up of ≥ 6 weeks was available in 289 cases (CoT 83, SuT 206 ). In this subset of patients relapse of infection occurred in six (7.2%) and 23 (11.2%) cases in the conservative and early surgical treatment group, respectively (p=0.69). Persisting neurological deficit was recorded in 21 (25.3%) of conservatively treated and 51 (24.8%) of surgically treated cases (p=0.92).
Conclusion: Whereas relapse rates and persisting neurological deficit were not found to differ significantly, the results of this international data analyses, with their respective limitations, clearly support the growing evidence of a significantly reduced mortality rate after surgical therapy for primary pyogenic spondylodiscitis when compared to conservative treatment regimen.

Within Europe, fundamentally different therapeutic concepts are found. Therefore, the aim of this study was to compare the outcome of patients who received surgical vs. antibiotic treatment alone for primary pyogenic spondylodiscitis in an international cohort analysis.
Methods: The retrospectively compiled databases of tertiary high-volume spine centers served as the baseline for this study. A follow-up of ≥ 6 weeks was available in 289 cases (CoT 83, SuT 206 ). In this subset of patients relapse of infection occurred in six (7.2%) and 23 (11.2%) cases in the conservative and early surgical treatment group, respectively (p=0.69). Persisting neurological deficit was recorded in 21 (25.3%) of conservatively treated and 51 (24.8%) of surgically treated cases (p=0.92).
Conclusions: Whereas relapse rates and persisting neurological deficit were not found to differ significantly, the results of this international data analyses, with their respective limitations, clearly support the growing evidence of a significantly reduced mortality rate after surgical therapy for primary pyogenic spondylodiscitis when compared to conservative treatment regimen.
Can right‐wing terrorism increase support for far‐right populist parties and if so, why? Exploiting quasi‐random variation between successful and failed attacks across German municipalities, we find that successful attacks lead to significant increases in the vote share for the right‐wing, populist Alternative für Deutschland (AfD) party in state elections. Investigating channels, we find that successful attacks lead to differential increases in turnout which are mainly captured by the AfD. Using the German SOEP, a longitudinal panel of individuals, we investigate terror’s impact on individual political attitudes. We first document that people residing in municipalities that experience successful or failed attacks are indistinguishable. We then show that successful terror leads individuals to prefer the AfD, adopt more populist attitudes and report significantly greater political participation at the local level. Terror also leads voters to migrate away from (some) mainstream parties to the AfD. We also find differential media reporting: successful attacks receive more media coverage among local and regional publishers, coverage which makes significantly more use of words related to Islam and terror. Our results hold despite the fact that most attacks are motivated by right‐wing causes and targeted against migrants. Moreover, successful attacks that receive the most media coverage have nearly double the effect on the AfD vote share in state elections and they also increase the AfD vote share in Federal elections, highlighting media salience as a driver of our overall results.
During the very well attended year congresses of EANS in Barcelona and EUROSPINE in Frankfurt we proudly awarded two authors for the Best Paper in the Brain Section respectively Spine Section of Brain and Spine in the past academic year (July 2022 - June 2023).

Mobilität ist eine wesentliche Voraussetzung, um an außerhäuslichen Aktivitäten teilzunehmen und somit am gesellschaftlichen Leben teilhaben zu können. Da das Nutzen von Verkehrsmitteln mit Kosten verbunden ist, kann dies insbesondere für Personen mit geringen Einkünften eine Mobilitätsbarriere darstellen und das Risiko erhöhen, von mobilitätsbezogener sozialer Exklusion betroffen zu sein.
Eine besondere und tragende Rolle fällt dem öffentlichen Personennahverkehr zu (ÖPNV), auf den wir mit diesem methodischen Beitrag eingehen möchten: Zunächst tragen wir zusammen, wie finanzielle ÖPNV-Erschwinglichkeit quantifiziert werden kann, wobei wir auf Grundlage internationaler Fachliteratur zwei Ansätze (beobachtete und potenzielle Erschwinglichkeit) unterscheiden. Anschließend diskutieren wir mögliche Indikatoren für die ÖPNV-Erschwinglichkeit, die auf dem Einkommen privater Haushalte basieren. Darüber hinaus skizzieren wir einen GIS-basierten Indikator für den Nutzen des Deutschlandtickets, das seit Mai 2023 bundesweit im ÖPNV gilt.
Mobilität ist eine Grundvoraussetzung, um am gesellschaftlichen Leben teilhaben zu können. Ist der Möglichkeitsraum potenzieller Ortsveränderungen eingeschränkt – beispielsweise durch räumliche, finanzielle oder körperliche Barrieren oder Ängste –, wird von Mobilitätsarmut gesprochen. Wie Mobilitätsarmut auf der Verwaltungsebene verhindert werden kann, zeigt dieser Policy Brief des Projektes Social2Mobility. Diese Handlungsempfehlungen basieren auf Erkenntnissen des Projektes Social2Mobility und adressieren insbesondere Verwaltungsmitarbeitende der Fachplanungen Verkehrs-, Raum- und Sozialplanung von der Kommunal- bis zur Landes- und Bundesebene. Ziel des Projektes Social2Mobility war es, die soziale Teilhabe armutsgefährdeter Personen durch eine Steigerung ihrer Mobilitätsoptionen zu stärken. Der Policy Brief erläutert die Problematik von Mobilitätsarmut, thematisiert deren Relevanz und zeigt verschiedene Facetten von Mobilitätsarmut auf. Er beinhaltet fünf verschiedene Handlungsfelder zur Verhinderung von Mobilitätsarmut. Der Policy Brief soll zu einer dezernats- und abteilungsübergreifenden Zusammenarbeit der Fachplanungen Sozial-, Raum- und Verkehrsplanung zur Lösung von Mobilitätsarmut anregen. Synergieeffekte und gegenseitige Potentiale bei einer Zusammenarbeit werden dabei herausgestellt. Hierzu gehören beispielsweise die Anwendung von Verkehrsnachfragemodellen in der Sozialplanung oder die Berücksichtigung vielfältiger unterschiedlicher Lebenslagen und der daraus hervorgehenden Mobilitätsbedarfe in der Verkehrsplanung.
Financial poverty, mobility and social participation are interrelated. This nexus makes old-age poverty a highly relevant issue in terms of transport-related social exclusion in an ageing society. To understand how financial poverty affects older people’s travel practices and how they cope with their limited financial resources, we conducted qualitative interviews with low-income older people (aged 60 and above) in Ronnenberg (Hanover region, Germany). Although all the respondents have comparably limited financial resources, using a practice theory perspective along the elements of materials, competences and meanings (Shove et al., 2012), we identified three different types of low-income older people by their travel practices: (i) active older people with multifaceted social interactions, (ii) neighbourhood-oriented older people with local interactions and (iii) home-centred older people with few social interactions. From our analysis, we conclude that financial poverty shapes each element of low-income older people’s travel practices and thereby increases the risk of transport-related social exclusion: (i) certain materials have to be financed, which is challenging or even impossible due to financial poverty; (ii) meanings of travel practices are strongly linked to other practices, thus, if (also for financial reasons) no or a limited range of destinations are mentioned, travel practices are restricted in their frequency and distance; and (iii) necessary competences to be mobile and engage in social activities despite low financial means seem to be differentially available or - more cautiously formulated – less often utilised by low-income older people, which may lead to them abandoning travel practices and related social activities.
Although exercise guidelines now recommend exercise for patients with MCI, the long-term effects of exercise in patients with MCI has not been reviewed systematically. The aim was to assess (1) the effectiveness of exercise and physical activity (EXPA) interventions in improving long-term patient-relevant cognitive and non-cognitive outcomes in people with mild cognitive impairment, (2) how well the included trials reported details of the intervention, and (3) the extent to which reported endpoints were in line with patient preferences that were assessed in patient workshops. Following PRISMA guidelines, we performed a systematic review and meta-analysis including randomized controlled trials. A total of ten studies were included after searching in six electronic sources from 1995 onwards. There is a trend that 6 + -month EXPA interventions improve global cognition 12 months after initiation. Evidence on long-term effects of EXPA interventions on non-cognitive health outcomes could not be meaningfully pooled and the individual studies reported mixed results. Workshop participants considered freedom from pain and stress, mood, motivation and self-efficacy to be important, but these outcomes were rarely addressed. Too little information is available on intervention details for EXPA programs to be replicated and confidently recommended for patients with MCI. Übermäßige Gewaltanwendungen durch Polizist:innen in Deutschland sind bislang nur in Ansätzen untersucht. Das Buch liefert umfassende wissenschaftliche Befunde zu einschlägigen Situationen und ihrer strafrechtlichen Aufarbeitung. Auf Basis einer Betroffenenbefragung mit über 3.300 Teilnehmenden und über 60 qualitativen Interviews stellen sich die Fälle als komplexe Interaktionsgeschehen dar, bei deren Aufarbeitung eine besondere Definitionsmacht der Polizei sichtbar wird.
In order to provide a reassessment of the Neotropical genus Pseudonannolene Silvestri, 1895, a cladistic analysis, biogeographic analysis, and taxonomic review were conducted in the present work. For the cladistic approach, 91 morphological characters were scored for 53 terminals as the ingroup and 10 as the outgroup. Three synapomorphies support the monophyly of the genus: presence of a longitudinal suture on the promentum, penial bases partially fused, and the internal branch of the gonopods surrounding the telopodite; and two homoplastic transformations: the lateral lobe of the collum densely striated and setae present up to the apical portion of the prefemoral process on the first leg-pair of males. The genus Pseudonannolene is recovered as sister-group of Epinannolene Brölemann, 1903 (Pseudonannoleninae). A total of 226 occurrence points were recorded for Pseudonannolene, with the majority of records from the Chacoan subregion, composed by Araucaria Forest, Atlantic, and Parana Forest provinces. The biogeographical searches using the Geographically explicit Event Model recovered two biogeographic reconstructions (cost of 79 000), with the vicariance events occurring more frequently in the deep clades, whereas sympatry and points of sympatry occurred in more inclusive clades. The first reconstruction recovered four vicariances, 13 sympatries, 4 points of sympatry, and 21 founder events, and the second reconstruction recovered four vicariances, 12–13 sympatries, 4–5 points of sympatry, and 21 founder events. The genus Pseudonannolene comprises 56 species, including 8 new species herein described: P. alata sp. nov., P. aurea sp. nov., P. bucculenta sp. nov., P. curvata sp. nov., P. granulata sp. nov., P. insularis sp. nov., P. morettii sp. nov., and P. nicolau sp. nov.; P. brevis Silvestri, 1902 and P. rugosetta Silvestri, 1897 are regarded as species inquirendae; a neotype of P. alegrensis Silvestri, 1897 is here proposed with male described for the first time. The following taxa are synonymized: P. canastra Gallo & Bichuette, 2020 and P. saguassu Iniesta & Ferreira, 2013 with P. ambuatinga Iniesta & Ferreira, 2013; P. marconii Iniesta & Ferreira, 2013 with P. longicornis (Porat, 1888); P. chaimowiczi Fontanetti, 1996, P. gogo Iniesta & Ferreira, 2013, P. rosineii Iniesta & Ferreira, 2014, P. taboa Iniesta & Ferreira, 2014, and P. longissima Iniesta & Ferreira, 2014 with P. microzoporus Mauriès, 1987; P. tricolor gracilis Brölemann, 1902 and P. tricolor rugosus Schubart, 1945 with P. tricolor Brölemann, 1902; P. auguralis Silvestri, 1902 with P. rocana Silvestri, 1902; and P. abbreviata Silvestri, 1902 with P. typica Silvestri, 1895. P. inops Brölemann, 1929 is proposed here as new status from P. bovei inops. A dichotomous identification key is presented to facilitate the species identification.
Cortical tracking of stimulus features (such as the envelope) is a crucial tractable neural mechanism, allowing us to investigate how we process continuous music. We here tested whether cortical and behavioural tracking of beat, typically related to rhythm processing, are modulated by pitch predictability. In two experiments (n=20, n=52), participants’ ability to tap along to the beat of musical sequences was measured for tonal (high pitch predictability) and atonal (low pitch predictability) music. In Experiment 1, we additionally measured participants’ EEG and analysed cortical tracking of the acoustic envelope and of pitch surprisal (using IDyOM). In both experiments, finger-tapping performance was better in the tonal than the atonal condition, indicating a positive effect of pitch predictability on behavioural rhythm processing. Neural data revealed that the acoustic envelope was tracked stronger while listening to atonal than tonal music, potentially reflecting listeners’ violated pitch expectations. Our findings show that cortical envelope tracking, beyond reflecting musical rhythm processing, is modulated by pitch predictability (as well as musical expertise and enjoyment). Stronger cortical surprisal tracking was linked to overall worse envelope tracking, and worse finger-tapping performance for atonal music. Specifically, the low pitch predictability in atonal music seems to draw attentional resources resulting in a reduced ability to follow the rhythm behaviourally. Overall, cortical envelope and surprisal tracking were differentially related to behaviour in tonal and atonal music, likely reflecting differential processing under conditions of high and low predictability. Taken together, our results show diverse effects of pitch predictability on musical rhythm processing.
Non-alcoholic steatohepatitis (NASH) and alcoholic steatohepatitis (ASH) are the leading causes of liver disease worldwide. To identify disease-specific pathomechanisms, we analyzed the lipidome, metabolome and immune cell recruitment in livers in both diseases. Mice harboring ASH or NASH had comparable disease severities regarding mortality rate, neurological behavior, expression of fibrosis marker and albumin levels. Lipid droplet size was higher in NASH than ASH and qualitative differences in the lipidome were mainly based on incorporation of diet-specific fatty acids into triglycerides, phosphatidylcholines and lysophosphatidylcholines. Metabolomic analysis showed downregulated nucleoside levels in both models. Here, the corresponding uremic metabolites were only upregulated in NASH suggesting stronger cellular senescence, which was supported by lower antioxidant levels in NASH as compared to ASH. While altered urea cycle metabolites suggest increased nitric oxide synthesis in both models, in ASH, this depended on increased L-homoarginine levels indicating a cardiovascular response mechanism. Interestingly, only in NASH were the levels of tryptophan and its anti-inflammatory metabolite kynurenine upregulated. Fittingly, high-content immunohistochemistry showed a decreased macrophage recruitment and an increased polarization towards M2-like macrophages in NASH. In conclusion, with comparable disease severity in both models, higher lipid storage, oxidative stress and tryptophan/kynurenine levels were seen in NASH, leading to distinct immune responses.
Unter den Bedingungen neoliberaler Meritokratie sind die Menschen ununterbrochen gefordert, sich selbst zu vermarkten, sich leistungsstark zu zeigen und ihre Position im Wettbewerb zu verbessern. Ins Zentrum des modernen Lebens rückt die Notwendigkeit, sich permanent zu thematisieren, zu optimieren und darzustellen. Die virtuellen sozialen Netzwerke sind in diesem Kontext als bedeutsame Formen der modernen Selbsttechnologie zu verstehen, in denen das kulturelle Begehren nach Authentizität und der Zwang zur Selbstdarstellung und Selbstverwirklichung einen Ausdruck finden. Die Frage nach dem Selbst oder genauer: die Frage, wie man zu diesem Selbst gelangt, stellt sich vor dem skizzierten Hintergrund auf mehrfache Weise. Das neoliberale Subjekt ist um eine ständige Sichtbarmachung des eigenen Selbst bemüht, das depressive Subjekt scheitert an diesem Imperativ und hat den Bezug zum eigenen Selbst verloren und das Subjekt der Psychiatrie versucht mithilfe von Expert*innen das entfremdete Selbst (wieder) zu erlangen. Innerhalb dieses Problemfeldes bewegt sich der vorliegende Beitrag. Am Beispiel des Schriftstellers Benjamin Maack soll der Frage nachgegangen werden, wie die Depression und die Erfahrung von Selbstverlust in einem Werk der Gegenwartsliteratur zur Sprache kommen. Im Gegenzug soll in einem zweiten Schritt untersucht werden, wie die Institution der psychiatrischen Klinik mit ihren Subjektivierungs- und Objektivierungsmechanismen im Text von Benjamin Maack dargestellt wird. Im Anschluss an Michel Foucault werden sowohl das literarische Schreiben als auch die psychiatrische Untersuchung als Geständnistechniken gedeutet. Die Entstehung der Literatur im modernen Sinne des Wortes wird in Beziehung gesetzt zu der Vervielfältigung und Ausbreitung der Geständnistechnologie im modernen abendländischen Staat. In diesem Beziehungsgeflecht, das es an späterer Stelle nachzuzeichnen gilt, nimmt das literarische Geständnis jedoch eine Sonderstellung ein und weicht von anderen Geständnistechniken ab.
Lebensqualität, kognitive Leistung und multisensorische Integrationsleistung bei NMOSD Patienten

Die Neuromyelitis-optica-Spektrum-Erkrankung (NMOSD) ist eine entzündliche Autoimmunerkrankung des zentralen Nervensystems (ZNS), die schubweise auftritt und meist in den Anfängen aufgrund der symptomatischen Ähnlichkeit mit der Multiplen Sklerose (MS) verwechselt wird. Primär manifestiert sich die NMOSD in Form von Sehstörungen und sensomotorische Lähmungserscheinungen. Im Krankheitsverlauf treten aber auch bei einem Großteil der Patienten kognitive Defizite auf, wobei vorwiegend das Gedächtnis, die Informationsverarbeitung und die Aufmerksamkeit betroffen sind, die nicht in Routineuntersuchungen erfasst werden. Kognitive Beeinträchtigungen wurden bereits bei der MS beschrieben. Ebenso spielt eine verminderte Lebensqualität bei beiden Erkrankungen eine große Rolle. Analog zu Untersuchungen bei MS Patienten, die gezeigt haben, dass kognitive Beeinträchtigungen mitunter ursächlich für die niedrige Lebensqualität sind, wird in dieser Arbeit postuliert, dass auch bei NMOSD Patienten das Ausmaß an kognitiven Dysfunktionen mit dem Grad an Einbußen in der Lebensqualität zusammenhängt. Ferner sollen weitere Prädiktoren ermittelt werden, welche einen Einfluss auf die Lebensqualität haben, wie bereits bestätigt körperliche Einschränkungen. Es wird erwartet, dass NMOSD Patienten von einer verminderten Lebensqualität berichten, die von den schlechteren Ergebnissen in den neuropsychologischen Tests vorhergesagt werden kann.
Zur Untersuchung der Kognition wurde in der vorliegenden Arbeit neben etablierten neuropsychologischen Tests auch die multisensorische Integrationsleistung mithilfe des SiFI Paradigmas angewandt, welche bereits bei MS Patienten und Patienten mit leichten kognitiven Defiziten (mild cognitive impairment; MCI) auffällige Daten lieferte und für eine Testung der globalen Kognitionsleistung genutzt werden konnte. Der Grund für den Einsatz der SiFI waren die nachgewiesenen Hirnkorrelate bei multisensorischer Integration, welche ebenfalls bereits bei kognitiver Dysfunktion festgestellt wurden, wie Atrophien, Konnektivitätsstörungen und Auffälligkeiten in der Transmission bestimmter Neurotransmitter. Ziel dieser Anwendung ist eine Implementierung der SiFI in den Klinikalltag zur erleichterten Erfassung kognitiver Defizite. Viele bekannte neuropsychologischen Tests sind entweder zu teuer, zu lang, abhängig von der sprachlichen Fähigkeit oder für die Patienten zu anstrengend. Die SiFI wäre daher eine gute Alternative als Marker kognitiver Defizite.
20 NMOSD Patienten wurden zu ihrer Lebensqualität (EQ-5D) sowie ihrem psychopathologischen Zustand (SCL-90-R) befragt und es wurde eine umfassende neuropsychologische Testung durchgeführt. Zur Diagnostik der multisensorischen Integrationsleistung wurde die SiFI Aufgabe herangezogen. Die Ergebnisse deuten auf eine verminderte kognitive Leistung mit mittelhohen Werten in den Fragebögen zur Lebensqualität. NMOSD Patienten nahmen die Illusion in der SiFI Aufgabe bei längeren Intervallen wahr, vergleichbar mit MS und MCI Patienten. Dies deutet auf eine verzögerte Integration sensorischer Informationen.
Angefangen mit einem Einblick über die Erkrankung und Darstellung des bisherigen Wissenschaftsstands zu den einzelnen Konstrukten und ihrer Zusammenhänge wird das Studiendesign vorgestellt und die Ergebnisse angegeben und interpretiert. Abschließend folgen eine kritische Beurteilung und Zusammenfassung der vorliegenden Daten mit Ausblick auf weitere Forschungsziele.
Mit "Corpus Delicti" zeigt Zeh eine biopolitische Kontrollmacht auf, die in der Fluchtlinie von Michel Foucaults und Giorgio Agambens biopolitischen Theorien auf dem menschlichen Körper fußt: Während jedoch Foucault die Einwirkung der Biomacht auf der sozio-politischen Ebene lokalisiert, nähert sich Giorgio Agamben dem biopolitischen Grundgedanken aus einer juristischen Perspektive an. "Corpus Delicti" absorbiert beide biopolitischen Annäherungen und veranschaulicht sie vor dem Hintergrund einer Gesundheitsdiktatur, in der die individuelle Freiheit der Gesundheit weichen muss; lieber gesund, schmerzfrei und lang lebend als frei. Mit diesem Ziel, allen Bürgern*innen die Gesundheit zu garantieren, verwaltet Zehs fiktiver Staat, die METHODE, totalitär die Körper seiner Bürger*innen und stößt dabei auf erstaunlich geringen Widerstand, denn die Angst vor Krankheit und Tod führt zur innerlichen Akzeptanz normierten Verhaltens im Namen der zur Staatsräson erhobenen Gesundheit. Mit diesem Beitrag möchte ich versuchen, Michel Foucaults und Giorgio Agambens Theoretisierung von Biopolitik so weit zu erklären, dass die biopolitischen Anknüpfungen in "Corpus Delicti" beleuchtet und veranschaulicht werden können. Vor dem Hintergrund des Verständnisses von Foucaults und Agambens biopolitischen Theorien und einer kurzen inhaltlichen Darstellung von "Corpus Delicti" wird es möglich sein, die literarische Verarbeitung biopolitischen Regierens in "Corpus Delicti" genauer aufzuzeigen, und dies sowohl aus einem sozio-politischen als auch juristischem Blickwinkel.
Im Rahmen des von der KfW Stiftung geförderten Artenschutzprojektes „Erhaltungskulturen von bedrohten Pflanzen in Hessen“ wurden in den Jahren 2014–2018 von 15 gefährdeten Pflanzenarten in verschiedenen Regionen Hessens Samen gesammelt und im Botanischen Garten der Stadt Frankfurt am Main vermehrt. In Zusammenarbeit mit Naturschutzbehörden, Forstämtern und Naturschutzverbänden wurden dokumentierte Wiederansiedlungsmaßnahmen oder Populationsstärkungen durchgeführt. Überzähliges Saatgut wurde im Botanischen Garten eingefroren und kann für zukünftige Artenschutzprojekte verwendet werden. Bei den 15 gefährdeten Arten handelt es sich um Allium strictum, Festuca albensis, Fumana procumbens, Iris spuria, Jurinea cyanoides, Mibora minima, Moenchia erecta, Nigella arvensis, Poa badensis, Scleranthus verticillatus, Sedum villosum, Spergula pentandra, Ventenata dubia, Veronica acinifolia und Vicia orobus. Alle diese Arten sind als Verantwortungsarten Bestandteil der hessischen Biodiversitätsstrategie. Im Jahr 2021, also drei Jahre nach dem Ende des Projektes, ergab eine Erfolgskontrolle folgendes Ergebnis: Von insgesamt 50 Wiederansiedlungsversuchen an 34 Orten sind immerhin 25 (50 %) erfolgreich. Weitere 5 (10 %) sind möglicherweise ebenfalls gelungen, jedoch sind die neu begründeten Populationen sehr klein. Eindeutig misslungen sind 20 (40 %). Die Gründe für das Misslingen sind meist klar erkennbar und werden thematisiert: Manchmal war der Zielort ungünstig gewählt, oder die verwendete Ansiedlungsmethode erwies sich als ungeeignet. Das Projekt konnte dazu beitragen, unser Wissen über Ökologie und Verbreitung sowie Gefährdung und Schutzbedarf dieser Verantwortungsarten teils erheblich zu vermehren. Damit leistet es auch einen Beitrag zum Schutz der bereits bestehenden Altpopulationen.
Unsere bisherige Kenntnis zur Situation von Crepis praemorsa (Abbiss-Pippau) war ungenügend und wurde im Rahmen eines Kooperationsprojektes mit dem Hessischen Landesamt für Naturschutz, Umwelt und Geologie (HLNUG) erheblich verbessert. Die Untersuchungen zeigen, dass es in Hessen offenbar nur noch acht Vorkommen dieser Art gibt: Drei sehr kleine Reliktpopulationen an der Bergstraße, eine im nordhessischen Bergland und vier in der Rhön, davon zwei sehr kleine. Keine der Populationen kann als groß bezeichnet werden, die Anzahl der Individuen liegt zwischen wenigen Exemplaren bis zum niedrigen dreistelligen Bereich. Lediglich eine Population erscheint aktuell ungefährdet. Fehlende Nutzung oder Pflege und in der Folge Verbrachung und Verbuschung sowie Lichtmangel durch Beschattung sind maßgebliche Gefährdungen. Ohne aktive Hilfsmaßnahmen werden vier dieser acht Populationen in naher Zukunft erlöschen.
Betrachtet man die Anzahl der in Hessen ausgestorbenen Pflanzenarten über den gesamten Zeitraum ab Beginn der floristischen Erforschung, so scheint ein Zusammenhang zwischen der geringeren Anzahl ausgestorbener Arten in den letzten Dekaden und den gleichzeitig vermehrt ausgewiesenen Naturschutzgebieten zu bestehen. Es stellt sich daher die Frage, ob es auch einen kausalen Zusammenhang gibt, oder ob andere Faktoren einen Einfluss auf das Ausmaß an ausgestorbenen Pflanzenarten haben. Für die Entwicklung der Aussterberate in den kommenden Jahrzehnten wird es nötig werden, alle Vorkommen der derzeit vom Aussterben bedrohten Pflanzenarten dahingehend zu überprüfen, ob ihr Bestand gesichert und erhalten werden kann. So kann dann die Frage beantwortet werden, ob es gelungen ist, das Aussterben von Pflanzenarten in Hessen zu stoppen oder lediglich zu verlangsamen.
In diesem Artikel und weiteren geplanten Folgen werden Ergänzungen, Nachträge, Erläuterungen und Korrekturen sowohl zum von der Taunusflora (Wittig & al. 2022) erfassten Gebiet (Vortaunus, Hoher Taunus, kammnaher Hintertaunus) als auch zum bisher nicht bearbeiteten kammfernen Bereich des Hintertaunus publiziert. Bei den bisherigen Ergänzungen und Nachträgen handelt es sich um die Ergebnisse der Nachbestimmung eines Belegs (Allium christophii), den Fund einer für das Gebiet bisher nicht genannten Art (Hylotelephium vulgare) sowie um Hinweise auf ältere Funde (Bolboschoenus maritimus, Polystichum lonchitis) und einen neueren Fund (Luronium natans). Weiterhin werden die wichtigsten Ergebnisse der Begehungen sechs kammferner Rasterfelder präsentiert (Rote-Liste- und sonstige seltene Arten). Als selten werden diejenigen Arten angesehen, die im bisherigen Kartierungsgebiet in maximal 5 % (= 26) der insgesamt 523 Rasterfelder gefunden wurden. Außerdem werden in der Taunusflora enthaltene Fehler korrigiert.
G-protein-coupled receptors (GPCRs) from the largest family of receptors in the human body. They contain seven transmembrane helices. There are roughly 800-900 GPCR genes expressed in humans encoded by 4-5% of the human genome. These receptors are the most important signal transducers and play a crucial role in cell physiology and pathology, by using various extracellular stimuli to start complex intracellular signaling. GPCRs interact with a wide variety of stimuli from small molecules (photons, ions, amines) to large molecules (peptides, small proteins), and trigger downstream cascade effects by interacting with G-proteins, GPCR kinases, and ß-arrestin. Because of their crucial roles in many cellular functions, GPCRs are the most important drug targets for the pharmaceutical industry. Approximately 30% of the clinically approved drugs available in the market are against GPCRs. In this work achieved successful expression and purification of GPCRs from class-C and class-A families. Combined with biochemical experiments, DNP-ssNMR, and molecular simulation helped to decipher the mechanism of crosstalk between the allosteric modulator, and the orthosteric binding sites of the peptide receptor. The main findings and major highlights of this dissertation are outlined in the following paragraphs.
The calcium-sensing receptor (CaSR) belongs to the GPCR class-C family and contains a large extracellular domain. This receptor regulates Ca2+ homeostasis in blood and its absorption in the kidney and bone. To understand the molecular and structural mechanisms of these receptors their cDNAs were cloned into the pPICZ and pOET1 vectors to express them in Pichia pastoris and in Sf9 insect cells respectively. The CaSR was successfully expressed heterologously in Pichia pastoris and in the insect cell with high yield. The purified receptor purified in LMNG shows no aggregation in a monomeric state. Further optimization was performed to use it for cryo-EM sample preparation and structure determination. In 2nd part of the thesis, different mini G (mini Gs, mini Gi, mini Gqs, and mini Gsi) DNA constructs were made and expressed in E. coli. It's challenging to obtain active GPCR structures due to the instability of G-protein or G-protein-bound receptors. In this work, all mini-G proteins and chimera mini-G-protein-maltose binding protein (MBP) were cloned and expressed in E. coli and purified with a His-trap column with high purity.
In the last part of the thesis, to decipher the mechanism of allosteric modulation of orthosteric binding sites in the bradykinin receptor was produced and characterized in insect cells. Angiotensin I converting enzyme inhibitors (ACEIs), are very important drugs and are widely used for the treatment of hypertension, congestive heart failure, and diabetic neuropathy. These drugs target primarily the catalytic zinc center of the ACE. It has been shown that enalaprilat, a well-known ACEI, binds to a proposed zinc-binding site on hB1R and even directly activates the receptor. To obtain information on the influence of ACEIs on the receptor-peptide complex, and to have a better understanding of the molecular mechanism and structural plasticity of the bradykinin receptor and PAM, we used the three commercially available ACEIs captopril, enalaprilat, and lisinopril for our studies. An important result of this thesis is that though enalaprilat, captopril, and lisinopril all have similar functional properties in humans, each one regulates the orthosteric binding site of hB1R in a unique way. These findings provide atomic insights into the allosteric modulation of the bradykinin receptor. This study along with the effects of ACEI on the binding sites of receptors also deciphers the effects of the Zn2+ as well as the crosstalk between zinc binding sites and ACEI compounds. The binding of allosteric modulators induces distinct endogenous binding, which might aid in creating new possibilities in the pharmaceutical field.
DNA binding redistributes activation domain ensemble and accessibility in pioneer factor Sox2

More than 1600 human transcription factors orchestrate the transcriptional machinery to control gene expression and cell fate. Their function is conveyed through intrinsically disordered regions (IDRs) containing activation or repression domains but lacking quantitative structural ensemble models prevents their mechanistic decoding. Here we integrate single-molecule FRET and NMR spectroscopy with molecular simulations showing that DNA binding can lead to complex changes in the IDR ensemble and accessibility. The C-terminal IDR of pioneer factor Sox2 is highly disordered but its conformational dynamics are guided by weak and dynamic charge interactions with the folded DNA binding domain. Both DNA and nucleosome binding induce major rearrangements in the IDR ensemble without affecting DNA binding affinity. Remarkably, interdomain interactions are redistributed in complex with DNA leading to variable exposure of two activation domains critical for transcription. Charged intramolecular interactions allowing for dynamic redistributions may be common in transcription factors and necessary for sensitive tuning of structural ensembles.
The interaction of Eph receptor tyrosine kinases with their transmembrane ligands; the ephrins, is important for the regulation of cell-cell communication. Ephrin-Eph signaling is probably best known for the discrimination of arterial and venous territories by repulsion of venous endothelial cells away from those with an arterial fate. Ultimately, cell repulsion is mediated by initiating the collapse of the actin cytoskeleton in membrane protrusions. Here, we investigated the role of the Ena/VASP family of actin binding proteins in endothelial cell repulsion initiated by ephrin ligands. Human endothelial cells dynamically extended sheet-like lamellipodia over ephrin-B2 coated surfaces. While lamellipodia of control siRNA transfected cells rapidly collapsed, resulting in a pronounced cell repulsion from the ephrin-B2 surfaces, the knockdown of Ena/VASP proteins impaired the cytoskeletal collapse of membrane protrusions and the cells no longer avoided the repulsive surfaces. Mechanistically, ephrin-B2 stimulation elicited the EphB-mediated tyrosine phosphorylation of VASP, which abrogated its interaction with the focal adhesion protein Zyxin. Nck2 was identified as a novel VASP binding protein, which only interacted with the tyrosine phosphorylated VASP protein. Nck links Eph-receptors to the actin cytoskeleton. Therefore, we hypothesize that Nck-Ena/VASP complex formation is required for actin reorganization and/or Eph receptor internalization downstream of ephrin-Eph interaction in endothelial cells, with implications for endothelial navigation and pathfinding.

Campanula cervicaria steht in Hessen kurz vor dem Aussterben und wurde im Rahmen des Kooperationsvertrages zwischen der Botanischen Vereinigung für Naturschutz in Hessen (BVNH) und dem Land Hessen untersucht. Das Ziel der Untersuchung war, herauszufinden wo Campanula cervicaria in Hessen noch vorkommt und wie groß die Populationen sind. In Hessen sind aktuell noch 20 Populationen vorhanden. Fast 90 % der Populationen befinden sich in der Umgebung von Lißberg, Hirzenhain und Gedern im Westlichen Unteren Vogelsberg. Zwei weitere liegen bei Jossa im Spessart und bei Ober-Gleen im Nördlichen Vogelsberg-Vorland. Die größte Population befindet sich an der Kreisstraße 216 bei Hirzenhain am Forsthaus Luisenlust. Campanula cervicaria wächst in Hessen an sonnenexponierten Wegböschungen und Windwurfflächen, meist zusammen mit Hypericum hirsutum und Brachypodium sylvaticum. Um auch zukünftig die Bestände von Campanula cervicaria zu sichern, müsste ein Artenhilfskonzept erarbeitet werden.
Der autobiographisch markierte Prosatext "The Argonauts" von Maggie Nelson aus dem Jahr 2015 kreist um die Beziehungsgeflechte der Ich-Erzählerin und zugleich Autorin Maggie Nelson, die sich in den queeren Künstler*innenszenen der US-amerikanischen Westküste bewegt. [...] Dieser Aufsatz geht der Frage nach, inwiefern "The Argonauts" als paradigmatisches Beispiel innovativer zeitgenössischer Literatur durch die Mittel der Autotheorie neue Weisen der Subjektivität erzeugt, welche verkörpert, hybrid und relational sind, und daher auch (bio-)politisch relevant gemacht werden können. Der Fokus auf die biopolitische Dimension bildet eine neue Perspektive auf Nelsons vielrezipierten Text, ebenso wie durch die Literaturanalyse ein neuer Blick auf aktuelle biopolitische Diskurse geschaffen werden kann. Zunächst soll der Begriff der Biopolitik mit Bezug auf Paul B. Preciado und Rosi Braidotti vor dem Hintergrund des globalisierten, digitalisierten 21. Jahrhunderts abgesteckt werden, in dessen Kontext auch "The Argonauts" eingebettet ist. [...] In einem nächsten Schritt soll auf die Verhandlung von queerer Identität und deren biopolitische Implikationen in "The Argonauts" genauer eingegangen werden, um des Weiteren das Verhältnis von Inhalt und Form unter die Lupe zu nehmen. Im Text lassen sich verschiedene Weisen der Transformation feststellen, die sowohl Nelsons Reflexionen um Sprache und Identität prägen als auch die Prämissen ihres experimentellen Schreibens bilden. Schließlich wird im letzten Abschnitt noch einmal der Begriff der Techno-Biopolitik mit den Ergebnissen der Analyse zusammengeführt.
Epithelial cells enable essential physiological functions, including absorption, morphogenesis, secretion, and transport. To execute these functions, epithelial cells often form three-dimensional shapes that include curved sheets of cells surrounding a pressurized fluid-filled lumen. These three-dimensional tissues (called domes) are essential for organ function, but when they are not working properly, developmental defects, inflammation, and cancer can ensue. Recently, it has been shown that the cells that form domes show active superelasticity on micropatterned plates.
We show here that the immortalized renal proximal tubule epithelial cell line, LLC-PK1, stereotypically forms tubules in 10 days. Tubule formation takes place in 4 stages. When cells are plated on a culture dish, they form a monolayer on the 1st day; on the 3rd day, three-dimensional structures are formed, called domes; and after the 4.5th day, these domes start fusing to begin the transition stage and transit to the tubule stage. At the end of the 10th day, differentiated, elongated, and matured tubes form (Figure 3.1). Therefore, tubule formation is a self-organized, stereotypic morphogenetic program under long-term, unperturbed tissue culture conditions.
We propose that tubulogenesis is a two-step process in proximal tubules by doming and wrapping. The process begins with dome formation, and as the cell layers come together in the transition stage at the edge of the dome, this leads to the formation of the lumen of the eventual tubule. We also found that F-actin provides the mechanical strength during the formation of these three-dimensional structures during tubule formation. To better understand this 4-step process on a molecular level, we performed proteomics of tubule formation to identify the different proteins that play a significant role in proximal tubule development. Importantly, we identified proximal tubule markers like synaptopondin, angiotensin 1-10, collectrin, polycystin 1, and polycystin 2. These proteins play an important role in renal tube formation and differentiation.
Cell division is carried out by highly conserved cyclin-CDK complexes, which phosphorylate various cellular components. Cyclin-CDKs act differently depending on the cell cycle phase and work cooperatively to create DNA replication and cytokinesis. Therefore, we identified that cyclin-B1, marker of proliferation Ki-67, the RAD51 recombinase, and proliferating cell nuclear antigen (PNCA) are upregulated in the monolayer stage, and the expression decreases as tubule formation takes place. The proximal tubule reabsorbs 60-65% of the glomerulus filtrate. Therefore, it requires a lot of energy generated by using the fatty acid oxidation (FAO) pathway. In our model, we found FAO expression is higher than that of the other metabolic pathways.
We found expression of an intricate protein network in mitochondria, which we interpret as a sign of mitochondrial homeostasis being vital for the FAO pathway to work. Furthermore, we also identified different types of transporters at each stage of proximal tubule formation, and we could recognize different cytoskeletal components playing a significant role in each stage of proximal tubule formation, for instance, at the monolayer stage, vimentin expression is high, and its expression is reduced as tubules form. Hence, this 2D system, at this step of characterization, seems suitable to use to study differential transport protein expression and how this might relate to physiological functions and syndromes.
Next, we inhibited different transporters using specific inhibitors and analyzed the effect on dome and tubule formation. We identified that Na+/K+ ATPase and vacuolar H+ ATPase play a significant role in the process of epithelial dynamics. Digoxin (a Na+/K+ ATPase inhibitor) treatment inhibits dome and tubule formation. Bafilomycin (a v-ATPase inhibitor) treatment demonstrated a delay in dome and tube formation. Therefore, this study shows that this 2D proximal tubule novel system can be used for screening of pharmacological leads in the context of specific aspects of kidney physiology.
Despite the recent success in growing kidney organoids, they are not well suited to investigate various pathophysiological conditions in vitro for several reasons: They grow in 3D and form a tissue that later needs to be dissected/cleared and stained to investigate pathophysiological changes. Moreover, organoids require complex and expensive protocols for generation and are challenging to use in screening approaches. Therefore, we set out to demonstrate feasibility for our 2D system using normal renal epithelial cells, which are the origin of various pathological conditions, to study pathophysiological conditions.
Hintergrund: Der Rettungsdienst versorgt täglich viele Patient/-innen in unterschiedlichen Umgebungen und ist damit auch potentieller Überträger nosokomialer Infektionen. Zur Händehygiene, als entscheidende Säule der Infektionsprophylaxe, liegen bislang nur wenige Daten aus dem Rettungsdienst vor.
Methoden: Prospektive multizentrische Studie mit Fragebogen zur Selbst- und Fremdeinschätzung der Compliance und beeinflussender Faktoren (abgeleitet von der WHO Perception Survey for Health-Care Workers) sowie direkte Compliance-Beobachtung nach WHO-Standard bei Rettungsdienstpersonal zweier Berufsfeuerwehren in Deutschland.
Ergebnisse: Es wurden 207 Fragebögen eingereicht und während ca. 66h Beobachtungszeit wurden 674 Händedesinfektionsgelegenheiten protokolliert. Der präventive Effekt der HH wurde allgemein von den Mitarbeitenden anerkannt. Die Selbsteinschätzung (MW: 80%) und beobachtete Compliance-Rate (38%) zeigten eine deutliche Diskrepanz und die Compliance variierte zwischen den verschiedenen Indikationen. Besonders niedrig zeigte sich die Compliance rund um die Durchführung aseptischer Tätigkeiten. Hier zeichnete sich ein geringes Risikobewusstsein für nicht sichtbare Verunreinigungen ab. Hürden für die Umsetzung der Händehygiene stellten vor allem die Vorrangigkeit anderer Maßnahmen, Unterbrechung des Arbeitsablaufes und Zeitmangel dar.
Schlussfolgerungen: Die beobachtete Compliance-Rate im Rettungsdienst lag unterhalb der innerklinischen Durchschnittswerte. Insbesondere die Compliance im Rahmen aseptischer Tätigkeiten muss dringend gesteigert werden. Dies erfordert einen multimodalen Lösungsansatz, der die Optimierung der Ausbildung, Algorithmen, Materialverfügbarkeit und Praktikabilität der Händedesinfektion im Rettungsdienst beinhaltet.
In natural environments, background noise can degrade the integrity of acoustic signals, posing a problem for animals that rely on their vocalizations for communication and navigation. A simple behavioral strategy to combat acoustic interference would be to restrict call emissions to periods of low-amplitude or no noise. Using audio playback and computational tools for the automated detection of over 2.5 million vocalizations from groups of freely vocalizing bats, we show that bats (Carollia perspicillata) can dynamically adapt the timing of their calls to avoid acoustic jamming in both predictably and unpredictably patterned noise. This study demonstrates that bats spontaneously seek out temporal windows of opportunity for vocalizing in acoustically crowded environments, providing a mechanism for efficient echolocation and communication in cluttered acoustic landscapes.
One Sentence Summary: Bats avoid acoustic interference by rapidly adjusting the timing of vocalizations to the temporal pattern of varying noise.
RNAs are key players in life as they connect the genetic code (DNA) with all cellular processes dominated by proteins. The dynamics study of RNA modifications has become an important part of epitranscriptomics field, as they are reversible and dynamically regulated far more than originally thought. Several evidences portrait a catalog of RNA modifications and their links to neurological disorders, cancers, and other diseases. Therefore, a deeper investigation of RNA modifications dynamics including their specific profile, biosynthesis, maturation and degradation is required for pioneering disease diagnostics and potential therapeutics development.
Mammalian tissues reveal diverse physiology and functions, despite sharing identical genomes and overlapping transcription profiles. So far, most research on this diversity were referred to variable transcriptomic processing among tissues and differential post-translational modifications that tune the activity of ubiquitous proteins to each tissue’s needs. However, study of epitranscriptome dynamics relevance to tissues’ functions is not yet revealed. There are a few reports on mouse RNA modification profiles, which are focused on only one type of RNA and limited types of modifications. The first part of my dissertation aims to generate a comprehensive tissue-specific as well as RNA species-specific investigation of all existing RNA modifications, as well as investigating potential codon as an effector of translation diversity among tissues. Using isotope dilution mass spectrometry, I created a library including absolute quantification of 24 tRNA modifications, and up to 22 rRNA modifications. I find an almost identical pattern of modifications in 28S- and 18S-rRNA subunits, but different levels of most modifications in 5.8S-rRNA or tRNA among highly metabolic active organs to e.g. heart or spleen. The findings suggest a high degree of similarity between quantities of modifications between presented data to all previous literature, confirming that it is a suitable model to study the tissue-based RNA modification patterns.
The most noticeable difference exhibited was tRNA modifications, which suggests a discerning tRNA engagement in translation between different organs. This can be a good start for investigation of codon bias in enriched genes of specific tRNA modifications among different tissues that may cause differential translation pattern, causing organs diversity. Moreover, 5.8S rRNA data showed an organ-specific pattern, which proposes functional diversity of this rRNA subunit among different organs. Future studies must investigate the possible implications of organ-specific 5.8S rRNA modifications functions, to elucidate the core of the observed variations.
Abundance of RNA modifications is carefully regulated in cells. Part of this regulation is achieved by activity of enzymes removing RNA modifications, named RNA erasers. Literature has provided proof of demethylation activity of AlkBH family on different types of RNA. For instance, AlkBH5 is known to remove m6A in mRNA, and both AlkBH3 and AlkBH1 are reported to demethylate m1A and m3C in tRNA. So far, RNA erasers are mainly studied in vitro and direct in vivo studies are missing.
Mass spectrometry is a promising approach in the identification and quantification of many RNA modifications. However, mass spectrometric analysis by nature, offers only a static view of nucleic acid modifications, and fails to account for their cellular dynamics. Nucleic Acid Isotope Labeling coupled Mass Spectrometry (NAIL-MS) was developed as a powerful technique which differentiates among remaining, co-transcriptional and post-transcriptional incorporation of a target RNA modification. This temporal resolution captures the dynamic nature of RNA modifications, and offers absolute and relative quantification of all existing nucleosides in any given RNA sequence, including different isotopologues and isotopomers.
The objective of this study was to uncover the first “direct” iv vivo data on AlkBH1, 3 and 5 activities in demethylating each of their specific substrates. I investigated the RNA modification changes through pulse-chase experiments in collaboration with my colleagues Dr. Kayla Borland and Dr. Felix Hagelskamp. A remarkable observation was that AlkBH3 protein -but not AlkBH1- was overexpressed under methylating reagent treatment in vivo. These findings suggest that AlkBH3 -but not AlkBH1- is a methylation damage induced enzyme, that potentially triggers ASCC-AlkBH3 alkylation repair complex after aberrant methylation damage by MMS treatment. However, using NAIL-MS method, we could not detect any significant effect on demethylation activity of the enzymes in tRNA, rRNA or mRNA towards the possible substrates m6A, m1A, m3C, m5C and m7G in vivo. These distinct outcomes can be partially explained by probable existence of other unidentified demethylases that compensate for AlkBHs demethylation activity; or more probably, demethylation may still arise by remaining active AlkBHs to restore the original levels of the observed RNA modifications, since a stronger KD or a complete knockout of AlkBHs genes was not possible. Further research on fully knocked out AlkBHs genes can provide stronger evidence on unidentified demethylation activities in HEK cells.
Neutron stars are unique laboratories for the investigation of the high density properties of bulk matter. In this work, the astrophysical constraints for a phase transition from hadronic matter to deconfined quark matter are examined thoroughly. A scheme for relating known astrophysical observables such as mass, radius and tidal deformability to the parameter space of such a transition is devised and applied to the set of data currently available.
In order to span a wide parameter space, a highly parameterizable relativistic mean field equation in compliance with chiral effective field theory results is used, where the stiffness of the equation of state can be varied via the effective mass at saturation density. The phase transitions are modelled using a Maxwell construction and assumed to be of first order, with a constant speed of sound quark matter model. The resulting equations of state are analyzed and divided into four categories, which can be used to constrain the parameter space that allows phase transition. It is highlighted, that a subset of this parameter space would even be detectable without the need of higher precision measurements. A phase transition at high densities is shown to be particularly promising in this regard. Finally, the groundwork is laid to apply the equation of state used in this work for supernova or merger simulations, by extending it to non-zero temperatures.
Der folgende Beitrag interessiert sich für das Verhältnis des biopolitischen Machtmodells zu Phänomenen moderner Serialisierung und möchte wahrnehmbar machen, in welch großem Ausmaß Technologien der Serialität für die Beobachtung, das Verständnis und die Manipulation bzw. Beherrschung des Lebens eingesetzt worden sind. Dabei soll deutlich werden, dass die Beziehungen zwischen Leben und Serialisierung derart umfangreich und ubiquitär sind, dass eine theoretische Beschreibung der Biopolitik ohne die Berücksichtigung des Konzepts 'Serialität' kaum sinnvoll möglich ist. Das mag teilweise auch daran liegen, dass beide Begriffe, also sowohl die 'Biopolitik' als auch die 'Serie' einen nur schwer zu beherrschenden Bedeutungsumfang aufweisen, der dazu verführt, allzu viele Phänomene mit diesen Konzepten zu beschreiben. Um dem etwas entgegenzuwirken, werden im Folgenden verschiedene Bereiche unterschieden, in denen das 'Leben' und die 'Serie' seit dem 19. Jahrhundert aufeinandertreffen, so dass biopolitische Serialisierung konkreter adressiert und eine allzu vage Verwendung der Begriffe vermieden werden kann. Diese Bereiche betreffen die Serialisierung der Wahrnehmung des Lebens, die Serialisierung der Lebensverhältnisse durch Mechanisierungsprozesse in modernen Industriegesellschaften und die massenmediale Serialisierung, die abschließend noch etwas mit Blick auf die periodische Presse und dem Beispiel des von 1923 bis 1935 erschienenen Magazins "Das Leben" konkretisiert wird.
This research article examines the dual impact of protests on COVID-19 spread, a challenge for policymakers balancing public health and the right to assemble. Using a game theoretical model, it shows that protests can shift infection risks between counties, creating a dilemma for regulators. The empirical study analyzes two German protests in November 2020 using proprietary data from a bus-shuttle service, finding evidence to support the assumption that protests can shift infection risks. The article concludes by discussing the implications of these findings for policymakers, highlighting that regulators’ individually rational strategic decisions may lead to inefficient outcomes.
Asymptotic giant branch (AGB) stars are responsible for the production of the main component of the solar s-process distribution. Despite enormous progress in the theoretical modeling of these objects over the last few decades, many uncertainties remain. The still-unknown mechanism leading to the production of 13C neutron source is one example. The nucleosynthetic signature of AGB stars can be examined in a number of stellar sources, from spectroscopic observations of intrinsic and extrinsic stars to the heavy-element isotopic composition of presolar grains found in meteorites. The wealth of available observational data allows for constraining the processes occurring in AGB interiors. In this view, we discuss recent results from new AGB models including the effects of mixing triggered by magnetic fields, and show comparisons of the related s-process nucleosynthesis with available observations.
WE PRESENT OUR VISION OF OMNISCIENTDB, A NOVEL DATABASE THAT LEVERAGES THE IMPLICITLY STORED KNOWLEDGE IN LARGE LANGUAGE MODELS TO AUGMENT DATA SETS FOR ANALYTICAL QUERIES OR MACHINE LEARNING TASKS. OMNISCIENTDB EMPOWERS USERS TO AUGMENT DATA SETS BY MEANS OF SIMPLE SQL QUERIES AND THUS HAS THE POTENTIAL TO DRAMATICALLY REDUCE THE MANUAL OVERHEAD ASSOCIATED WITH DATA INTEGRATION. IT USES AUTOMATIC PROMPT ENGINEERING TO CONSTRUCT APPROPRIATE PROMPTS FOR GIVEN SQL QUERIES AND PASSES THEM TO A LARGE LANGUAGE MODEL LIKE GPT-3 TO CONTRIBUTE ADDITIONAL DATA, AUGMENTING THE EXPLICITLY STORED DATA. OUR INITIAL EVALUATION DEMONSTRATES THE GENERAL FEASIBILITY OF OUR VISION, EXPLORES DIFFERENT PROMPTING TECHNIQUES IN GREATER DETAIL, AND POINTS TOWARDS FUTURE RESEARCH.

Memory consolidation tends to be less robust in childhood than adulthood. However, little is known about the corresponding functional differences in the developing brain that may underlie age-related differences in retention of memories over time. This study examined system-level memory consolidation of object-scene associations after learning (immediate delay), one night of sleep (short delay), as well as two weeks (long delay) in 5-to-7-year-old children (n = 49) and in young adults (n = 39), as a reference group with mature consolidation systems. Particularly, we characterized how functional neural activation and reinstatement of neural patterns change over time, assessed by functional magnetic resonance imaging combined with representational (dis)similarity analysis (RSA). Our results showed that memory consolidation in children was less robust (i.e., more forgetting) compared to young adults. For correctly retained remote memories, young adults showed increased neural activation from short to long delay in neocortical (parietal, prefrontal and occipital) and cerebellar brain regions, while children showed increased neural activation in prefrontal and decrease in neural activity in parietal brain regions over time. In addition, there was an overall attenuated scene-specific memory reinstatement of neural patterns in children compared to young adults. At the same time, we observed category-based reinstatement in medial-temporal, neocortical (prefrontal and parietal), and cerebellar brain regions only in children. Taken together, 5-to-7-year-old children, compared to young adults, show less robust memory consolidation, possibly due to difficulties in engaging in differentiated neural reinstatement in neocortical mnemonic regions during retrieval of remote memories, coupled with relying more on gist-like, category-based neural reinstatement.
Human functional brain connectivity can be temporally decomposed into states of high and low cofluctuation, defined as coactivation of brain regions over time. Rare states of particularly high cofluctuation have been shown to reflect fundamentals of intrinsic functional network architecture and to be highly subject-specific. However, it is unclear whether such network-defining states also contribute to individual variations in cognitive abilities – which strongly rely on the interactions among distributed brain regions. By introducing CMEP, a new eigenvector-based prediction framework, we show that as few as 16 temporally separated time frames (< 1.5% of 10min resting-state fMRI) can significantly predict individual differences in intelligence (N = 263, p < .001). Against previous expectations, individual’s network-defining time frames of particularly high cofluctuation do not predict intelligence. Multiple functional brain networks contribute to the prediction, and all results replicate in an independent sample (N = 831). Our results suggest that although fundamentals of person-specific functional connectomes can be derived from few time frames of highest connectivity, temporally distributed information is necessary to extract information about cognitive abilities. This information is not restricted to specific connectivity states, like network-defining high-cofluctuation states, but rather reflected across the entire length of the brain connectivity time series.
This research introduces a new 3D bioprinter that incorporates live imaging of the bioprinted tissue with high resolution and high-speed capabilities. The printer employs a light sheet-based system to photocrosslink polymers into hydrogels at a printing speed of up to 0.66 mm³/s with a resolution of 15.7 µm. A significant advancement of this bioprinter is its ability to track cells and bioink during crosslinking, which enables real- time evaluation of the 3D-bioprinted structure’s quality. Fibroblast cells were encapsulated using this method, and the viability was evaluated directly after bioprinting and seven days after encapsulation, which was found to be high (83% ± 4.34%). Furthermore, a full- thickness skin construct was bioprinted and maintained in culture for 6 weeks, demonstrating the long-term viability and physiological relevance of the bioprinted tissue. The usage of solid-state laser beam scanning devices could enhance bioprinting’s speed and precision. This fast and accurate light-based bioprinter offers a promising platform for generating customizable 3D-printed structures with viable long-term cultures.
"The Eyes of God run over all the world" : Biopolitik in Margaret Atwoods "The Handmaid's Tale"

Im Folgenden soll untersucht werden, wie sich eine Biopolitik in "The Handmaid's Tale" gestaltet. So sollen die Kontrollmaßnahmen der Republik Gilead betrachtet werden sowie die dabei wichtige Rolle der Religion. Die Auswirkungen dieser Maßnahmen auf die Bevölkerung werden vor allem an der Protagonistin und Erzählerin 'Offred' sichtbar, da sie ihre Perspektive auf das Leben in Gilead zeigt. Vor allem an ihrer Person werden Möglichkeiten des Widerstandes deutlich, obwohl sie als Handmaid - Gileads 'Variante' einer Leihmutter - einer der hierarchisch niedrigsten Stellungen in Gilead angehört.
Im Wesentlichen liefern die beiden Romane - dies soll im Folgenden gezeigt werden - Beispiele für eine Kontinuitätsthese, die Martin Weiß als Lesart benennt: die Fortführung bzw. das unsichtbare (Wieder-)Aufgehen souveräner Macht im biopolitischen Regulieren und Disziplinieren. Um die 'verbundene Polarität' plastisch werden zu lassen, wähle ich die Metapher des Gängelbands als Veranschaulichung, die eine doppelte Lesart in sich vereint: Zum einen steht sie für einen lebenssichernden, biopolitischen Eingriff, der trotz Kontrolle durch die charakteristische 'Steigerung des Lebens' ein repressives Movens ausschließt; zum anderen erweist sie sich als gleichsam rückläufiges Modell, das das 'alte Recht' der autoritären Zucht rehabilitiert. In diesem historischen Rahmen "gesellschaftlicher Transformationsprozesse", manifestiert sich ein (anti-)biopolitisches Wechselspiel. Dieses Spannungsverhältnis findet sich als literarische Reflexion nicht nur in Grosses Roman, sondern auch in Goethes "Wilhelm Meisters Lehrjahre" (1795/1796). Die Protagonisten beider Romane treffen auf sektenähnliche Gruppen, die sie wie am Gängelband führen und so direkt in das Leben der 'Jünglinge' eingreifen. Wie sich dies genau ausgestaltet, soll sich in der folgenden Analyse zeigen.
Dual coding theories of knowledge suggest that meaning is represented in the brain by a double code, which comprises language-derived representations in the Anterior Temporal Lobe and sensory-derived representations in perceptual and motor regions. This approach predicts that concrete semantic features should activate both codes, whereas abstract features rely exclusively on the linguistic code. Using magnetoencephalography (MEG), we adopted a temporally resolved multiple regression approach to identify the contribution of abstract and concrete semantic predictors to the underlying brain signal. Results evidenced early involvement of anterior-temporal and inferior-frontal brain areas in both abstract and concrete semantic information encoding. At later stages, occipito-temporal regions showed greater responses to concrete compared to abstract features. The present findings shed new light on the temporal dynamics of abstract and concrete semantic representations in the brain and suggest that the concreteness of words processed first with a transmodal/linguistic code, housed in frontotemporal brain systems, and only after with an imagistic/sensorimotor code in perceptual and motor regions.
This paper analyses, if the Intertemporal Guarantee of Freedom, that was developed by the German Federal Constitutional Court (GFCC), can be used to expand the protection of human rights against the harms of climate change. The case of the Swiss Senior Women shows that there are jurisdictions, where the Intertemporal Guarantee of Freedom could be applied to improve standing and the control standard of states’ climate change action. Within international law bodies with jurisdiction over human rights treaties there are distinctive standards of protection against the harms of climate change. A major deficit within the international human rights protection against climate change lies within the focus on the positive obligations and the corresponding wide margin of appreciation granted to the states. The Intertemporal Guarantee of Freedom could provide a protection expansion in this regard, especially in the case of the European Court of Human Rights. It could also enable and legitimise present human rights concerns focused on the future actions of states following their past inaction. One considerable hurdle that is not addressed by it are procedural hurdles like the Plaumann formula applied by the European Court of Justice. The Intertemporal Guarantee of Freedom cannot solve major problems for climate change litigation like procedural hurdles. Yet, it can provide a new approach for complaints to address unambitious mitigation legislation which will lead to future human rights infringements.
Im Nachwort von "Dune" schreibt Brian Herbert, der Sohn von Frank Herbert, über den Krieg gegen die Maschinen im Dune-Universum. In diesem Krieg befreiten sich die Menschen von den intelligenten Maschinen und es wurde ein Gesetz zum Verbot der Herstellung der Maschinen aufgestellt: "Thou shall not make a machine in the likeness of a human mind". Aber, und das ist die Tragik dieses Ausgangspunktes, es entsteht im Gegensatz zum Kampf um die eigene Souveränität eine neue Beherrschung des Menschen. Diese Wende, welche die Künstliche Intelligenz aus der Welt verbannt hat, aber die Biopolitik mit anderen Techniken fortführt, soll im Folgenden anhand der Bereiche Religion, Politik und Psychologie herausgearbeitet werden. Denn indem in "Dune" die künstliche Intelligenz verbannt wird, treten Herrschafts- und Kontrolltechniken wieder in den Vordergrund, die in den aristokratisch-feudalen Gesellschaften eine bedeutende Rolle einnahmen.
In dem Roman "Never Let Me Go" von Kazuo Ishiguro erzählt Kathy H. kurz vor Ablauf ihrer Lebenszeit in einer Rückblende ihre Lebensgeschichte. Ihr Rückblick beginnt in Hailsham, einem Internat, wo sie zusammen mit Tommy und Ruth ihre Kindheit erlebt. Sie erfährt recht früh, dass sie und alle Kinder, die das Internat besuchen, Klone sind und eines Tages der Gesellschaft, von der sie geschaffen wurden, ihr Leben bzw. ihre Körper in Form einer Organspende bereitstellen müssen. [...] Die Klone sind in den Augen der Nicht-Klone weniger wert, als diejenigen, denen sie nachgebildet sind und werden von den Nicht-Klonen durch verschiedene Mechanismen biopolitisch manipuliert und gesteuert. Kathys und Tommys Bestreben ihre Liebe zu leben und die Suche der Protagonist*innen nach der eigenen Identität stehen im Fokus der Handlung. Deshalb stelle ich mit dem Titel "Klon, Person, Identifikation" die Frage nach der Menschlichkeit der Protagonist*innen an den Anfang meiner Überlegungen zu dem Roman von Kazuo Ishiguro. Im weiteren Verlauf meines Aufsatzes beschäftige ich mich mit der Frage nach den biopolitischen Zusammenhängen zwischen der Identitätsfindung auf der einen Seite und der Identitätsentwicklung der Klone auf der anderen Seite.
Die Paulinenpflege Winnenden e.V. wurde 1823 gegründet als Rettungshaus für verwahrloste Kinder. Bald wurde daraus eine Doppelanstalt einerseits für Kinder, die aus sozialen Gründen nicht zuhause leben konnten, andererseits für hör- und sprachbehinderte Kinder. Zum 200-jährigen Jubiläum im Jahr 2023 erscheint dieses wissenschaftliche Werk. Dieses Werk erscheint zusätzlich in weitgehend identischer Weise als Buch.
The traditional view on coding in the cortex is that populations of neurons primarily convey stimulus information through the spike count. However, given the speed of sensory processing, it has been hypothesized that sensory encoding may rely on the spike-timing relationships among neurons. Here, we use a recently developed method based on Optimal Transport Theory called SpikeShip to study the encoding of natural movies by high-dimensional ensembles of neurons in visual cortex. SpikeShip is a generic measure of dissimilarity between spike train patterns based on the relative spike-timing relations among all neurons and with computational complexity similar to the spike count. We compared spike-count and spike-timing codes in up to N > 8000 neurons from six visual areas during natural video presentations. Using SpikeShip, we show that temporal spiking sequences convey substantially more information about natural movies than population spike-count vectors when the neural population size is larger than about 200 neurons. Remarkably, encoding through temporal sequences did not show representational drift both within and between blocks. By contrast, population firing rates showed better coding performance when there were few active neurons. Furthermore, the population firing rate showed memory across frames and formed a continuous trajectory across time. In contrast to temporal spiking sequences, population firing rates exhibited substantial drift across repetitions and between blocks. These findings suggest that spike counts and temporal sequences constitute two different coding schemes with distinct information about natural movies.
Die dystopische Jugendliteratur hat sich in den letzten Jahren zu einem politischen, kulturellen und ästhetischen Genrephänomen entwickelt. [...] Die anhaltende Bedeutung der dystopischen Literatur in der Populärkultur zeigt bereits auf, dass sich auch aus literaturwissenschaftlicher Perspektive mit diesem Genrephänomen auseinandergesetzt werden sollte. Im aktuellen Forschungsdiskurs der Literaturwissenschaften wird der Aktualität und Relevanz dieses Phänomens bereits begegnet, indem sowohl das Genre als auch einzelne Werke auf verschiedene Aspekte hin untersucht werden. Die Forschungsschwerpunkte liegen hierbei jedoch vordergründig in der Untersuchung von psychologischen und soziologischen Aspekten der Protagonist*innen im Hinblick auf Fragen der Ökokritik, der Gender Studies oder der Kulturwissenschaften. Jedoch fehlen im gegenwärtigen Diskurs fast gänzlich Fragen nach einer eventuell vorherrschenden Biopolitik im Genre oder aber in konkreten Werken, obwohl diese in zahlreichen der fiktiven Weltbilder eindeutig gegeben ist. Dieser Beitrag erweitert den bereits bestehenden Diskurs um die Fragestellung, welche Formen einer dystopischen Biopolitik im zeitgenössischen Jugendroman am Beispiel der Reihe "Divergent" zu finden sind und wie diese literaturwissenschaftlich analysiert werden können. Da eine vollständige Analyse der drei Romane und all ihrer relevanten Aspekte zur Biopolitik den Rahmen dieses Beitrags überschreiten würde, werden einige Analyse-Schwerpunkte gebildet. Diese bestehen sowohl aus der Untersuchung des Wirklichkeitsmodells der Romane und der Thematik Biopolitik als auch aus der literarischen Darstellung. Durch die Untersuchung dieser Aspekte wird deutlich, inwiefern literarische Werke, auch dystopische Romanreihen, als Aushandlungs- und Reflexionsraum für aktuelle biopolitische Fragestellungen fungieren können. Zunächst werden die vier elementaren biopolitischen Inhalte der Romane herausgearbeitet und in Bezug zu den Ausführungen Foucaults gesetzt: Gesellschaftssystem der verschiedenen Fraktionen, neue technische Möglichkeiten der fremdgesteuerten Bewusstseinskontrolle, das Leben als und in einem Experiment und die Differenzierung von genetisch perfekten und genetisch defekten Menschen. Anschließend wird ein Blick auf die literarische Umsetzung in Form der Raum-/ Zeitstruktur und der Charakterkonstellation geworfen. Diese beiden Aspekte unterstützen das bewusste Einfühlen der Leser*innenschaft in die Romansituation und ermöglichen auf diese Weise sowohl eine Auseinandersetzung mit der Biopolitik aus ethischer Sicht als auch eine Vergegenwärtigung der Parallelen zur Alltagswelt der Leser*innenschaft. Abschließend werden ein Fazit und ein kurzer Ausblick formuliert.

ONCE THEY RECEIVE INFORMATION ON THIS EROSION CHANNEL, THEY UPDATE PERCEPTIONS AND EXPECTATIONS ABOUT OWN NET NOMINAL POSITIONS. LEARNING ABOUT THE INFLATION-INDUCED EROSION OF NOMINAL POSITIONS CAUSALLY AFFECTS CHOICES IN HYPOTHETICAL REAL-ESTATE TRANSACTIONS AND ACTUAL CONSUMPTION. THE FINDINGS SUGGEST THAT HOUSEHOLD WEALTH MEDIATES THE SENSITIVITY OF CONSUMPTION TO INFLATION ONCE HOUSEHOLDS ARE AWARE OF THE BALANCE-SHEET EFFECTS OF INFLATION.
Nok Eisen : zentralnigerianische Eisenverhüttung in der Mitte des ersten Jahrtausends vor Christus

Based on excavations, excavation documentation and archaeometallurgical analyses, this thesis aims to characterise Nok iron production in central Nigeria through a contextually based investigation.
In 2010, 2011, 2013 and 2016, the Nok research project at Goethe University Frankfurt/Main in collaboration with the National Commission for Museums and Monuments, Nigeria excavated 27 iron-smelting furnaces from 8 sites 60 kilometres north of Abuja. All furnaces date around the middle of the first millennium BCE. Absolute dates, relative pottery chronology and terracotta figurine finds in furnace contexts suggest their affiliation to the Nok context. In comparison, all 27 furnaces resemble each other closely regarding their design and spacial arrangement. The numbers of furnaces per site, furnace width, furnace wall angle and thickness as well as pits beneath the furnaces are just some features with similar qualities. The similarities of the smelting sites also extend into their finds: the structure of tuyères and their position in situ as well as macroscopic slag morphology and distribution. Find morphology and distribution as well as furnace structure suggest a highly standardized way of Nok iron production. However, archaeometallurgical analyses show heterogeneous use of raw materials between sites and/or furnaces. In similarly structured furnaces different kinds of iron ore were smelted leaving a high iron content in the respective slags. This hints at an early stage of iron production in which the smelting process was limited to one operative set-up.
We investigate the development of the directed, v1, and elliptic flow, v2, in heavy ion collisions in mid-central Au+Au reactions at Elab=1.23A GeV. We demonstrate that the elliptic flow of hot and dense matter is initially positive (v2>0) due to the early pressure gradient. This positive v2 transfers its momentum to the spectators, which leads to the creation of the directed flow v1. In turn, the spectator shadowing of the in-plane expansion leads to a preferred decoupling of hadrons in the out-of-plane direction and results in a negative v2 for the observable final state hadrons. We propose a measurement of v1−v2 flow correlations and of the elliptic flow of dileptons as methods to pin down this evolution pattern. The elliptic flow of the dileptons allows then to determine the early-state EoS more precisely, because it avoids the strong modifications of the momentum distribution due to shadowing seen in the protons. This opens the unique opportunity for the HADES and CBM collaborations to measure the Equation-of-State directly at 2-3 times nuclear saturation density.
Chloritoid and kyanite coexist in metapelites from the high-pressure/low-temperature Massa Unit in the Alpi Apuane metamorphic complex (Northern Apennines, Italy). The composition of chloritoid is extremely variable throughout the Massa Unit. Fe-chloritoid occurs in association with hematite-free, graphite-bearing schists, whereas strongly zoned Fe-Mg chloritoid is found with hematite and kyanite. We investigated the effect of different bulk Fe2O3 contents in controlling chloritoid composition through phase equilibria modelling of four selected samples, representative of the different chloritoid-bearing parageneses found in the Massa Unit. The ferric iron content, measured through wet chemical titration, ranges from 0 (graphite-chloritoid schist) to 73% of the total iron (hematite-chloritoid schist). We show that Mg-rich chloritoid compositions and stability of kyanite at greenschist to blueschist facies conditions can be reproduced in the MnO–Na2O–K2O–FeO–MgO–Al2O3–SiO2–H2O–TiO2–O (MnNKFMASHTO) chemical system only considering the presence of significant amounts of ferric iron as part of the bulk composition. The stabilization of kyanite at lower grade is directly linked to the presence of Fe2O3, which renders the reactive bulk rock composition effectively enriched in Al2O3 with respect to Fe and Mg. We also document that high Fe2O3 contents exacerbate the effect of chloritoid fractionation, producing strongly zoned Fe-Mg-chloritoid grains. Finally, the P–T modelling of the Massa Units performed in this study allows, for the first time, the recognition of a two-stage evolution at peak conditions, with an earlier pressure peak (1.2–1.3 GPa at 350–400°C), and a later thermal peak (0.7–1.1 GPa at 440–480°C), compatible with subduction, underthrusting and exhumation of the Adria continental margin during growth of the Northern Apennine orogenic wedge.
The oxidation state of sulfur in slab fluids is controversial, with both dominantly oxidized and reduced species proposed. Here we use in situ X-ray absorption spectroscopy analysis of sulfur-in-apatite to monitor changes in the oxidation state of sulfur during high-P metasomatism by slab fluids in the subduction channel. Our samples include a 73 cm continuous transect of reaction zones between a metagabbroic eclogite block and serpentinite matrix from a mélange zone on the island of Syros, Greece. The block core consists of garnet, omphacite, phengite, paragonite, epidote-clinozoisite, and rutile. In this region, apatite is only observed as elongate inclusions in omphacite cores. From the core outwards micas are increasingly replaced by epidote-clinozoisite, garnets are smaller and more frequent, pyrite + bornite is observed as inclusions in recrystallized omphacite, and apatite is increasingly abundant in the matrix and inclusions in garnet. A major transition at 48 cm separates an assemblage of Ca-Na amphibole, omphacite, chlorite, pyrite, and apatite from the inner garnet-bearing eclogite assemblages. Omphacite disappears from the assemblage at ~56 cm and amphibole compositions sharply transition to tremolite at 59 cm. Finally, the assemblage tremolite + talc + pyrite is observed after ~70 cm.Apatites in the eclogite assemblages exclusively display S6+ peaks in their absorption spectra. This includes apatite inclusions in omphacite in the least altered lithology, as well as matrix apatite and isolated apatite inclusions in garnet in the outermost metasomatized eclogite zone. In the intermediate pyrite-rich (~1-5 vol %) amphibole + omphacite + chlorite zone, apatite displays a strong S1- absorption peak in most grains, with rare analyses showing mixed S1- and S6+. Finally, apatite in the outermost tremolite-bearing assemblages only displays a S6+ peak. The pyrite-rich zone at 48 cm occurs at the initial interface between the serpentinite matrix and eclogite block, characterized by a dramatic decrease in Na content and Mg#. Our data suggest that reduction of S6+ in infiltrating fluids to S1- in pyrite became focused as Fe diffused across the steep Mg# gradient, resulting in pyrite precipitation. In contrast, S reduction in the Mg-rich tremolite-dominant portions of the transect was limited by a lack of Fe, resulting in low modes of pyrite and fluid buffered S6+ in apatite. Finally, S6+-bearing apatite is also observed in reaction zone lithologies from elsewhere on Syros, suggesting our observations are not isolated.Two important conclusions are drawn from these data and observations: (1) In the case of Syros, slab fluids at eclogite-facies conditions carried oxidized S6+, and (2) The interaction of these fluids with eclogites composed of ferrous-Fe silicates resulted in extensive sulfide precipitation.
RNA research is very important since RNA molecules are involved in various gene regulatory mechanisms as well as pathways of cell physiology and disease development.1 RNAs have evolved from being considered as carriers of genetic information from DNA to proteins, with the three major types of RNA involved in protein synthesis, including messenger RNA (mRNA), transfer RNA (tRNA), and ribosomal RNA (rRNA).2 In addition to the RNAs involved in protein synthesis numerous regulatory non-coding RNAs (ncRNAs) have been discovered in the transcriptome. The regulatory ncRNAs are classified into small ncRNAs (sncRNAs) with transcripts less than 200 nucleotides (nt) and long non-coding RNAs (lncRNAs) with more than 200 nt.3
LncRNAs represent the most diverse and versatile class of ncRNAs that can regulate cellular functions of chromatin modification, transcription, and post-transcription through multiple mechanisms.4 They are involved in the formation of RNA:protein, RNA:RNA and RNA:DNA complexes as part of their gene regulatory mechanism.4,5 The RNA:DNA interactions can be divided into RNA:DNA heteroduplex formation, also called R-loops, and RNA:DNA:DNA triplex formation. In triplex formation, RNA binds to the major groove of double-stranded DNA through Hoogsteen or reverse Hoogsteen hydrogen bonding, resulting in parallel or anti-parallel triplexes, respectively. In vitro studies have confirmed the formation of RNA:DNA:DNA triplexes.6 However, the extent to which these interactions occur in cells and their effects on cellular function are still not understood, which is why these structures are so exciting to study (Chapter I RNA:DNA:DNA Triplexes).
This cumulative thesis investigates several functional and regulatory important RNAs. The first project involves the improved biochemical and biophysical characterization of RNA:DNA:DNA triplex formation between lncRNAs of interest and their target genes. Triplex formation was confirmed by a series of experiments including electromobility shift assays (EMSA), thermal melting assays, circular dichroism (CD), and liquid state nuclear magnetic resonance (NMR) spectroscopy. The following is a summary of the main findings of these publications.
In research article 5.1, the oxygen-sensitive HIF1α-AS1 was identified as a functionally important triplex-forming lncRNA in human endothelial cells using a combination of bioinformatics techniques, RNA/DNA pulldown, and biophysical experiments. Through RNA:DNA:DNA triplex formation, endogenous HIF1α-AS1 decreases the expression of several genes, including EPH receptor A2 (EPHA2) and adrenomedullin (ADM), by acting as an adaptor for the repressive human silencing hub (HUSH) complex, which has been studied by our collaborators in the groups of Leisegang and Brandes.
2) Triplex formation between HIF1α-AS1 and the target genes EPHA2 and ADM was investigated in biochemical and biophysical studies. The EMSA results indicated that HIF1α-AS1 forms a low mobility RNA:DNA:DNA triplex complex with the EPHA2 DNA target sequence. The CD spectrum of the triplex showed distinct features compared to the EPHA2 DNA duplex and the RNA:DNA heteroduplex. Melting curve analysis revealed a biphasic melting transition for triplexes, with a first melting point corresponding to the dissociation of the RNA strand with melting of the Hoogsteen hydrogen bonds. The second, higher melting temperature corresponds to the melting of stronger Watson-Crick base pairing. Stabilized triplexes were formed using an intramolecular EPHA2 DNA duplex hairpin construct in which both DNA strands were attached to a 5 nucleotide (nt) thymidine linker. This approach allowed improved triplex formation with lower RNA equivalents and higher melting temperatures. By NMR spectroscopy, the triplex characteristic signals were observed in the 1H NMR spectrum, the imino signals in a spectral region between 9 and 12 ppm resulting from the Hoogsteen base pairing. To elucidate the structural and sequence specific Hoogsteen base pairs 2D 1H,1H-NOESY measurements of the EPHA2 DNA duplex and the HIF1α-AS1:EPHA2 triplex were performed. The 1H,1H-NOESY spectrum of the HIF1α-AS1:EPHA2 triplex with a 10-fold excess of RNA was semi-quantitatively analyzed for changes in the DNA duplex spectrum. We discovered, strong and moderate attenuation of cross peak intensities in the imino region of the NOESY spectrum. This attenuation was proposed to result from weakening of Watson-Crick base pairing by Hoogsteen hydrogen bonding induced by RNA binding. The Hoogsteen interactions can be mapped based on the analysis of the cross peak attenuation in the NOESY spectra, which we used to generate a structural model of the RNA:DNA:DNA triplex. These biophysical results support the physiological function of HIF1α as a triplex-forming lncRNA that recruits the HUSH-epigenetic silencing complex to specific target genes such as EPHA2 and ADM, thereby silencing their gene expression through RNA:DNA:DNA triplex formation.
Die labyrinthische Organisation des Erzählraums, die bisweilen dem Prinzip der Assoziation durch Josef K. zu folgen scheint, korrespondiert historisch mit der desorientierten Wirklichkeitserfahrung der Moderne - einem "Gefühl der 'Ohnmacht' und der 'Entfremdung' des 'modernen Menschen'". Nicht ohne Grund wird "Der Proceß" so in der verbreiteten sozialgeschichtlichen (und vor allem sozialkritischen) Lesart zur literarischen "Gestaltung der verwalteten Welt, der vielfältigen Einengungen und Bedrohungen, die das Individuum heute durch anonyme Mächte erfährt, zur Anklage des Kapitalismus, zur prophetischen Vorwegnahme der totalitären Regime des 20. Jahrhunderts oder, aktueller und à la Foucault, zur Verbildlichung der das Begehren unterdrückenden 'Macht'". Der vorliegende Beitrag unternimmt den Versuch, diese sozialgeschichtliche Lesart unter Hinzunahme des in der Kafka-Forschung bislang nur randläufig berücksichtigten Paradigmas der 'Biopolitik' bzw. 'Biomacht', wie es insbesondere von Michel Foucault analysiert wurde, noch einmal produktiv zu erweitern. [...] Franz Kafkas literarisches Werk befasst sich zweifellos immer wieder mit modernen Machtverhältnissen und Normierungsprozessen sowie auch mit den Individuen, die vor der (scheinbaren) Wahl stehen, sich zu assimilieren oder von eben jenen verschüttet zu werden. Dieser Beitrag wird erstens zu zeigen versuchen, wie das Gericht in Der Proceß durch seine Raumordnung als Metapher für eine breitere Machtökonomie der diegetischen Gesellschaftsordnung lesbar wird. Zweitens wird er der Frage nachgehen, inwieweit die Kategorie des Angeklagten als Schnittpunkt gelesen werden kann, der zwar intradiegetisch einen eigenen institutionellen und epistemologischen Stellenwert hat, auf der Metaebene allerdings der Logik des modernen Zusammenspiels bio- und disziplinarpolitischer Interventionen und Prozesse nachspürt.
Nach dem Anthropologen Kaushik Sunder Rajan bedeutet der Biokapitalismus die Überdeterminierung der Wirtschaft bei biopolitischen Entscheidungen. Die Überdeterminierung heißt in diesem Fall eine starke wirtschaftlich-politische Beeinflussung der Richtung wissenschaftlicher Forderung und Entwicklung. Ein extremes Beispiel für solch ein System ist in der Dystopie "Brave New World" (1932) vorzufinden, mit der sich dieser Aufsatz beschäftigt. Zuerst werden die Begriffe der Biopolitik nach Michel Foucault und des Biokapitalismus nach Kaushik Sunder Rajan definiert. Darauf aufbauend untersucht die Arbeit den Biokapitalismus innerhalb der Gesellschaft von Aldous Huxleys Roman. Dafür wird zunächst der Autor, sein zeitlicher Kontext und der Aufbau der dystopischen Gesellschaft thematisiert. Im Hauptteil befasst sich der Aufsatz mit folgender Fragestellung: Wie zeigt sich der Biokapitalismus in Huxleys "Brave New World"? Dabei sind die Ziele des Aufsatzes die Beantwortung der Fragestellung und die Herauskristallisierung der Gefahren dieser dystopischen Gesellschaft für unsere Gegenwart. Diese werden zum Schluss im Fazit zusammengefasst erläutert.
Transient receptor potential (TRP) ion channels are among the most well-studied classes of temperature-sensing molecules. Yet, the molecular mechanism and thermodynamic basis for the temperature sensitivity of TRP channels remains to this day poorly understood. One hypothesis is that the temperature-sensing mechanism can simply be described by a difference in heat capacity between the closed and open channel states. While such a two-state model may be simplistic it nonetheless has descriptive value, in the sense that it can be used to to compare overall temperature sensitivity between different channels and mutants. Here, we introduce a mathematical framework based on the two-state model to reliably extract temperature-dependent thermodynamic potentials and heat capacities from measurements of equilibrium constants at different temperatures. Our framework is implemented in an open-source data analysis package that provides a straightforward way to fit both linear and nonlinear van ‘t Hoff plots, thus avoiding some of the previous, potentially erroneous, assumptions when extracting thermodynamic variables from TRP channel electrophysiology data.

Was verstehen wir unter "Freigabe"? Das Freigeben, das Freigegebenwerden. Synonyme: Entlassung, Freilassung. [...] In Lois Lowrys 1993 erschienenem Roman "Hüter der Erinnerung" - im Original "The Giver" - ist die Tragweite dieses Terminus enorm: "Freigegeben zu werden ist für einen aktiven Bürger eine endgültige Sache, eine schreckliche Strafe, ein niederschmetternder Beweis menschlichen Versagens." Während es im Original "release" heißt, enthält die deutsche Übersetzung durch "frei" im Begriff im Kontext des Verfahrens eine Mehrdeutigkeit, die am Ende erörtert wird. Es soll im Folgenden sowohl mit dem Original als auch mit der Übersetzung textnah gearbeitet werden. [...] Es wird deutlich, dass mit dem Begriff des Freigebens eine ganz neue Denotation einhergeht, sodass "Freigeben" letztlich nichts anderes als ein Euphemismus ist, der die eigentliche Bedeutung, die Konsequenz des Vollzugs für die Sprecher*innen verschleiert. Ein ausgeprägtes Sprachregime und eine Vernichtungspolitik treten hier eng verflochten auf. Das Bezeichnen von Dingen, Handlungen und Ritualen ist dabei eine Voraussetzung für die Umsetzung des totalitären Regimes, wie im Folgenden unter anderem deutlich werden soll. Zusätzlich soll beispielhaft betrachtet werden, welche stilistischen Mittel für die Darstellung eines extremen biopolitischen Gesellschaftsmodells in einem Jugendroman für ein Lesepublikum ab dem jungen Alter von zwölf Jahren vorliegen. Dabei soll die Frage beantwortet werden, wie in "Hüter der Erinnerung" ein totalitäres System etabliert und welche Wirkung auf die Leser*innenschaft erzeugt wird. Lässt sich vielleicht eine gewisse schonende Vermittlung gegenüber der jungen Zielgruppe bezüglich des ernsten, 'belastenden' Themas feststellen?
Control of cell proliferation is critical for the lymphocyte life cycle. However, little is known on how stage-specific alterations in cell-cycle behavior drive proliferation dynamics during T-cell development. Here, we employed in vivo dual-nucleoside pulse labeling combined with determination of DNA replication over time as well as fluorescent ubiquitination-based cell-cycle indicator mice to establish a quantitative high-resolution map of cell-cycle kinetics of thymocytes. We developed an agent-based mathematical model of T-cell developmental dynamics. To generate the capacity for proliferative bursts, cell-cycle acceleration followed a 'stretch model', characterized by simultaneous and proportional contraction of both G1 and S phase. Analysis of cell-cycle phase dynamics during regeneration showed tailored adjustments of cell-cycle phase dynamics. Taken together, our results highlight intrathymic cell-cycle regulation as an adjustable system to maintain physiologic tissue homeostasis and foster our understanding of dysregulation of the T-cell developmental program.
Alzheimer’s Disease (AD) is a progressive and irreversible neurodegenerative disorder, characterized by the accumulation of abeta-amyloid aggregates, which triggers tau hyperphosphorylation and neuronal loss. While the precise mechanisms underlying neurodegeneration in AD are not entirely understood, it is known that loss of proteostasis is implicated in this process. Maintaining neuronal proteostasis requires proper transfer RNA (tRNA) modifications, which are crucial for optimal translation. However, research into tRNA epitranscriptome in AD is limited, and it is not yet clear how alterations in tRNA modifying enzymes and tRNA modifications might contribute to disease progression. Here, we report that expression of the tRNA modifying enzyme ELP3 is reduced in the brain of AD patients and amyloid AD mouse models, suggesting ELP3 is implicated in proteostasis dysregulation observed in AD. To investigate the role of ELP3 specifically in neuronal proteostasis impairments in the context of amyloid pathology, we analyzed SH-SY5Y neuronal cells carrying the amyloidogenic Swedish familial AD mutation in the APP gene (SH-SWE) or the wild-type gene (SH-WT). Similarly to the amyloid mouse models, SH-SWE exhibited reduced levels of ELP3 which was associated with tRNA hypomodifications and reduced abundance, as well as proteostasis impairments. Furthermore, the knock-down of ELP3 in SH-WT recapitulated the proteostasis impairments observed in SH-SWE cells. Importantly, the correction of tRNA deficits due to ELP3 reduction rescued and reverted proteostasis impairments of SH-SWE and SH-WT knock-down for ELP3, respectively. Additionally, SH-WT exposed to the secretome of SH-SWE or synthetic amyloid aggregates recapitulate the SH-SWE phenotype, characterized by reduced ELP3 expression, tRNA hypomodification and increased protein aggregation. Taken together, our data suggest that amyloid pathology dysregulates neuronal proteostasis through the reduction of ELP3 and tRNA modifications. This study highlights the modulation of tRNA modifications as a potential therapeutic avenue to restore neuronal proteostasis in AD and preserve neuronal function.
Endothelial tip cells are essential for VEGF-induced angiogenesis, but underlying mechanisms are elusive. Endothelial-specific deletion of EVL, a member of the mammalian Ena/VASP protein family, reduced the expression of the tip cell marker protein endothelial cell specific molecule-1 (Esm1) and compromised the radial sprouting of the vascular plexus in the postnatal mouse retina. The latter effects could at least partly be attributed to reduced VEGF receptor 2 (VEGFR2) internalization and signaling but the underlying mechanisms(s) are not fully understood. In the present study, we revealed that the expression of the long non-coding RNA H19 was significantly reduced in endothelial cells from postnatal EVL-/- mice and in siRNA-transfected human endothelial cells under hypoxic conditions. H19 was recently shown to promote VEGF expression and bioavailability via Esm1 and hypoxia inducible factor 1α (HIF-1α). Similar to EVL-/- mice, the radial outgrowth of the vascular plexus was significantly delayed in the postnatal retina of H19-/- mice. In summary, our data suggests that loss of EVL not only impairs VEGFR2 internalition and downstream signaling, but also impairs VEGF expression and bioavailability in the hypoxic retina via downregulation of lncRNA H19.
Vergesellschaftung von Salzpflanzen an Straßenrändern unter besonderer Berücksichtigung des Taunus

Im Taunus wurden an Straßen bisher fünf Arten festgestellt, die allgemein als Charakterarten der mitteleuropäischen Salzvegetation gelten und daher als Salzpflanzen im engeren Sinne bezeichnet werden können: Cochlearia danica, Plantago coronopus, Puccinellia distans, Spergularia marina und Trifolium fragiferum. Mit Ausnahme der Vorkommen von Trifolium fragiferum handelt es sich dabei oft um nur wenige weitere Arten enthaltende Dominanzbestände, wobei Trittpflanzen am häufigsten vertreten sind, so dass Straßenrandvegetation mit Puccinellia distans in der vorliegenden Literatur allgemein den Trittpflanzengesellschaften zugeordnet wurde (Plantaginetea majoris beziehungsweise Polygono-Poetea annuae). Entsprechendes gilt auch für viele Vorkommen von Plantago coronopus. Wie die vorliegende Arbeit zeigt, können aber einige der aus dem Taunus vorliegenden Aufnahmen mit Plantago coronopus zu den Salzwiesen im weiteren Sinne gestellt werden (Juncetea maritimi). Entsprechendes gilt für die hier präsentierte Vegetationsaufnahme eines Cochlearia-danica-Bestandes. Trifolium fragiferum wurde dagegen in einer als Flutrasen (Molinio-Arrhenatheretea, Agrostietalia stoloniferae) zu bezeichnenden Pflanzengesellschaft angetroffen.
To understand the neural mechanisms underlying brain function, neuroscientists aim to quantify causal interactions between neurons, for instance by perturbing the activity of neuron A and measuring the effect on neuron B. Recently, manipulating neuron activity using light-sensitive opsins, optogenetics, has increased the specificity of neural perturbation. However, using widefield optogenetic interventions, multiple neurons are usually perturbed, producing a confound -- any of the stimulated neurons can have affected the postsynaptic neuron making it challenging to discern which neurons produced the causal effect. Here, we show how such confounds produce large biases in interpretations. We explain how confounding can be reduced by combining instrumental variables (IV) and difference in differences (DiD) techniques from econometrics. Combined, these methods can estimate (causal) effective connectivity by exploiting the weak, approximately random signal resulting from the interaction between stimulation and the absolute refractory period of the neuron. In simulated neural networks, we find that estimates using ideas from IV and DiD outperform naive techniques suggesting that methods from causal inference can be useful to disentangle neural interactions in the brain.
Background Vasoplegic syndrome is frequently observed during cardiac surgery and resembles a complication of high mortality and morbidity. There is a clinical need for therapy and prevention of vasoplegic syndrome during complex cardiac surgical procedures. Therefore, we investigated different strategies in a porcine model of vasoplegia.
Methods We evaluated new medical therapies and prophylaxis to avoid vasoplegic syndrome in a porcine model. After induction of anesthesia, cardiopulmonary bypass was established through median sternotomy and central cannulation. Prolonged aortic cross-clamping (120 min) simulated a complex surgical procedure. The influence of sevoflurane-guided anesthesia (sevoflurane group) and the administration of glibenclamide (glibenclamide group) were compared to a control group, which received standard anesthesia using propofol. Online hemodynamic assessment was performed using PiCCO® measurements. In addition, blood and tissue samples were taken to evaluate hemodynamic effects and the degree of inflammatory response.
Results Glibenclamide was able to break through early vasoplegic syndrome by raising the blood pressure and systemic vascular resistance as well as less need of norepinephrine doses. Sevoflurane reduced the occurrence of the vasoplegic syndrome in the mean of stable blood pressure and less need of norepinephrine doses.
Conclusion Glibenclamide could serve as a potent drug to reduce effects of vasoplegic syndrome. Sevoflurane anesthesia during cardiopulmonary bypass shows less occurrence of vasoplegic syndrome and therefore could be used to prevent it in high-risk patients.

Beim Spielen kommt es offenbar aufs Maß an und auf die Umstände. Nicht oder nur schlecht spielen zu können gilt als Schwäche; umgekehrt erscheint es als riskant oder gefährlich, zu viel zu spielen, sich in Spielereien zu verlieren oder sogar ein falsches Spiel zu treiben. In der positiven Vorstellung des maßvollen, regelbewussten Spielens wirken bis heute Grundzüge anthropologischer Selbstbeschreibungen des 18. Jahrhunderts nach. In dieser Zeit rückte der Spielbegriff in den Fokus neuer ästhetischer Theorien, bevor er sich im 19. Jahrhundert als Gegenkonzept zu Ernst und Arbeit weiterentwickelte. Das Spiel wurde zum Aushandlungsort bürgerlichen Selbstverständnisses und gesellschaftlicher Regeln, zum Gegenstand von Theorie und Literatur.
All-optical closed-loop voltage clamp for precise control of muscles and neurons in live animals

Excitable cells can be stimulated or inhibited by optogenetics. Since optogenetic actuation regimes are often static, neurons and circuits can quickly adapt, allowing perturbation, but not true control. Hence, we established an optogenetic voltage-clamp (OVC). The voltage-indicator QuasAr2 provides information for fast, closed-loop optical feedback to the bidirectional optogenetic actuator BiPOLES. Voltage-dependent fluorescence is held within tight margins, thus clamping the cell to distinct potentials. We established the OVC in muscles and neurons of Caenorhabditis elegans, and transferred it to rat hippocampal neurons in slice culture. Fluorescence signals were calibrated to electrically measured potentials, and wavelengths to currents, enabling to determine optical I/V-relationships. The OVC reports on homeostatically altered cellular physiology in mutants and on Ca2+-channel properties, and can dynamically clamp spiking in C. elegans. Combining non-invasive imaging with control capabilities of electrophysiology, the OVC facilitates high-throughput, contact-less electrophysiology in individual cells and paves the way for true optogenetic control in behaving animals.
Bereits mit dem Titel dieses Bandes wird in Aussicht gestellt, darin "Biopolitik(en) in Literatur, Film und Serie" zu thematisieren. Mit der Überschrift dieses Beitrags wird hierzu analog angekündigt, die "Modi der Aushandlung menschlichen Lebens in (Gegenwarts-)Literatur, Film und Serie" zu untersuchen. Was jedoch meint 'in' im gegebenen Kontext? Worin besteht die Konnexion zwischen biopolitischer Thematik und künstlerischem Medium, die über die Präposition zum Ausdruck gebracht wird? Zunächst kann ein Auftreten biopolitischer Themen in Erzähltexten auf Ebene der 'histoire' festgestellt werden, insofern Biopolitik in ihren diversen Spielarten die Settings und Plots literarischer wie filmischer erzählender Texte ausgestaltet. [...] Weitergehend kann eine Wirksamkeit biopolitischer Fragestellungen in Erzähltexten auf Ebene des 'discours' konstatiert werden, sobald Biopolitik zum Reflexionsanlass eines erzählenden Textes wird. Über Szenario und Handlung hinausgehend, eröffnen semantische, narratologische oder metafiktionale Auseinandersetzungen mit biopolitischen Fragen neue Aushandlungs- und Reflexionsräume, in denen Biopolitik perspektiviert und problematisiert wird und mit denen eine kritische Revision durch die Rezipierenden initiiert wird. In dieser Variante biopolitischer Thematik in erzählenden Texten wird somit Biopolitik nicht nur zur Darstellung gebracht, sondern zur Disposition gestellt. In dieser Weise kann ein Erzähltext selbst als Beitrag zum biopolitischen Diskurs fungieren und als solcher Lebenswissen generieren. Auf diesen Prämissen aufbauend werden im Folgenden zunächst der Begriff der Biopoethik und die Besonderheiten biopoethischer Modi skizziert, bevor eine ausführlichere Betrachtung vier solcher Modi an konkreten Erzähltextbeispielen erfolgt. Als komplementäre Ergänzung zur Produktivität biopolitischer Diskurse, wie sie im voranstehenden Beitrag erarbeitet wird, soll so die Produktivität narrativer Biopolitik(en) verdeutlicht werden.
Wie fragil der scheinbare Differenzmarker der Emotionalität zwischen Mensch und Maschine ist, führt insbesondere das Genre des Science-Fiction-Films vor, das als ein Experimentierraum fungiert, in dem die biopolitischen Diskurse durch ästhetische bzw. poetische Konter-Diskurse aufgebrochen und hinterfragt werden. Diese Konterdiskursivität führt der Science-Fiction-Film "Ex Machina" (2015) eindrücklich vor, indem dieser anhand der als weiblich gegenderten und künstlich konstruierten AI und Protagonistin Ava (Alicia Vikander), auf der Ebene der Handlung die Frage verhandelt, ob die Grenze zwischen Mensch und Android teilweise aufgehoben werden kann, und damit verbunden die Frage, ob der vermeintliche Differenzmarker der Emotionalität nicht von Androiden produktiv gewendet werden könne, insofern er programmatisch in die entsprechenden Algorithmen eingebaut wird. Diese leitenden Überlegungen führen zur These des vorliegenden Beitrages: Die Protagonistin Ava bedient sich gegenüber den männlichen Figuren des Films der Strategie der Täuschung, indem Ava ihre künstlich angeeignete Emotionalität nutzt, um aus der Forschungsstation zu fliehen. Emotionalität kann entsprechend nicht (mehr) als eine Essenz des Menschen bzw. des Menschseins gelesen werden, sondern muss als ein elementares Konstrukt verstanden werden, das von der Maschine gegenüber den Menschen genutzt wird oder zumindest genutzt werden kann, um diese zu überlisten. Die vermeintliche moralische Überlegenheit des Menschen entpuppt sich folglich als ein grundlegender, menschlicher Denkfehler. Überlegen ist der Mensch gegenüber der Maschine nicht durch seine Emotionalität, vielmehr ist das Gegenteil der Fall: Weil der Mensch emotional ist, gelingt es der Maschine, ihn zu täuschen und sich somit als dem Menschen überlegen zu etablieren. Dergestalt führt "Ex Machina" explizit eine doppelte Grenzüberschreitung vor, indem der Film zum einen darauf aufbaut, dass Androiden scheinbar emotional handeln, da Avas Verhalten einen hoch rationalen Akt darstellt, der die künstlich erschaffene Frau gegenüber den Männern, Nathan (Oscar Isaac) und Caleb (Domhnall Gleeson), die sie beständig konstruieren, optimieren und testen, letztlich als überlegen präsentiert. Zum anderen führt "Ex Machina" anhand der Maschine Ava auf der Ebene des Gendering eindrücklich vor Augen, dass auch die männliche Überlegenheit und die damit einhergehende Geschlechterhierarchie ein reines Konstrukt ist. Der Homosozialität der beiden männlichen Figuren des Films steht die weiblich gegenderte Maschinenfrau Ava konträr gegenüber, auch wenn sie zunächst in diese integriert zu sein scheint, so dass die Frage nach der Repräsentation von Geschlecht und die nach den Verhandlungen zwischen den Geschlechtern nachdrücklich gestellt wird. Die damit aufgerufene Engführung von Männlichkeit und Überlegenheit sowie Weiblichkeit und Emotionalität wird im Verlauf des Films dezidiert aufgebrochen, sodass die Frage nach der Transgression gleich auf zweifache Weise gestellt wird, wenn man den Blick auf Ava richtet: 1. Inwiefern wird das Verhältnis zwischen Mensch und Maschine, in Bezug auf 'bios', an der weiblichen Androide Ava verhandelt? 2. Inwiefern erlaubt es Ava, Geschlechterverhältnisse neu zu denken, respektive das Verhältnis der Geschlechter, wie es im Film vorgeführt wird, einer neuen Betrachtung zu unterziehen? Um diese Fragen zu beantworten, werde ich in einem ersten Schritt die Differenz zwischen Mensch und Maschine, wie sie in "Ex Machina" inszeniert wird, in den Fokus der Betrachtungen stellen, um danach zu fragen, inwiefern das Verhältnis zwischen den 'menschlichen Männern' und der 'künstlichen Androide' das Konzept des 'bios' verhandelt. In einem anschließenden zweiten Schritt werde ich mich auf das Verhältnis zwischen den männlichen Figuren Caleb und Nathan sowie deren Beziehung zu Ava konzentrieren, um herauszuarbeiten, welche Geschlechterpolitik(en) in "Ex Machina" inszeniert wird, respektive werden.
The Asian tiger mosquito Aedes albopictus is currently spreading across Europe, facilitated by climate change and global transportation. It is a vector of arboviruses causing human diseases such as chikungunya, dengue hemorrhagic fever and Zika fever. For the majority of these diseases, no vaccines or therapeutics are available. Options for the control of Ae. albopictus are limited by European regulations introduced to protect biodiversity by restricting or phasing out the use of pesticides, genetically modified organisms (GMOs) or products of genome editing. Alternative solutions are thus urgently needed to avoid a future scenario in which Europe faces a choice between prioritizing human health or biodiversity when it comes to Aedes-vectored pathogens. To ensure regulatory compliance and public acceptance, these solutions should preferably not be based on chemicals or GMOs and must be cost-efficient and specific. The present review aims to synthesize available evidence on RNAi-based mosquito vector control and its potential for application in the European Union. The recent literature has identified some potential target sites in Ae. albopictus and formulations for delivery. However, we found little information concerning non-target effects on the environment or human health, on social aspects, regulatory frameworks, or on management perspectives. We propose optimal designs for RNAi-based vector control tools against Ae. albopictus (target product profiles), discuss their efficacy and reflect on potential risks to environmental health and the importance of societal aspects. The roadmap from design to application will provide readers with a comprehensive perspective on the application of emerging RNAi-based vector control tools for the suppression of Ae. albopictus populations with special focus on Europe.
In the recent past, we are making huge progress in the field of Artificial Intelligence. Since the rise of neural networks, astonishing new frontiers are continuously being discovered. The development is so fast that overall no major technical limits are in sight. Hence, digitization has expanded from the base of academia and industry to such an extent that it is prevalent in the politics, mass media and even popular arts. The DFG-funded project Specialized Information Service for Biodiversity Research and the BMBF-funded project Linked Open Tafsir can be placed exactly in that overall development. Both projects aim to build an intelligent, up-to-date, modern research infrastructure on biodiversity and theological studies for scholars researching in these respective fields of historical science. Starting from digitized German and Arabic historical literature containing so far unavailable valuable knowledge on biodiversity and theological studies, at its core, our dissertation targets to incorporate state-of-the-art Machine Learning methods for analyzing natural language texts of low-resource languages and enabling foundational Natural Language Processing tasks on them, such as Sentence Boundary Detection, Named Entity Recognition, and Topic Modeling. This ultimately leads to paving the way for new scientific discoveries in the historical disciplines of natural science and humanities. By enriching the landscape of historical low-resource languages with valuable annotation data, our work becomes part of the greater movement of digitizing the society, thus allowing people to focus on things which really matter in science and industry.
Bei über der Hälfte der Patienten mit einer operationsbedürftigen MI liegt gleichzeitig eine Insuffizienz der TK vor. In den meisten Fällen handelt es sich hierbei um eine funktionelle Insuffizienz welche aus einer RV-Pathologie, bedingt durch eine Volumen- oder Druckbelastung resultiert. In der Vergangenheit bestand die Überzeugung eine konservative Behandlung der TI sei ausreichend und nach Beheben der ursächlichen Grunderkrankung sei diese selbstlimitierend. Neuere Studien konnten jedoch belegen, dass eine bestehende TI auch nach operativer Versorgung einer ursächlichen LV-pathologie keine ausreichende Rückbildungstendenz aufweist, sondern im Verlauf sogar noch progredient ist. Die persistierende TI führt zu einer deutlich erhöhten Morbidität und Mortalität. Ist eine zweizeitige Operation an der TK im Verlauf erforderlich, so ist die Früh- und Spätmortalität deutlich erhöht und die Langzeitergebnisse sind schlecht. Diese Ergebnisse haben dazu geführt, dass die Indikation zur operativen Versorgung der TI ≥ 2 zu einem früheren Zeitpunkt und weniger restriktiv gestellt wird. Jedoch weisen die aktuellen ESC/EACTS-Guidelines insbesondere hinsichtlich der Versorgung einer TI < 2 nur einen niedrigen Evidenz-Grad auf, der richtige Operationszeitpunkt bleibt weiterhin umstritten. Der Grund für eine eher zurückhaltende Einstellung ist vor allem die Angst vor einer erhöhten Morbidität und Mortalität durch den additiven Eingriff. Zudem sind kaum Studien zu einer operativen Versorgung einer TI < 2 vorhanden. Ziel der vorliegenden Arbeit war daher der Gewinn weiterer Erkenntnisse, dieses bis dato kaum untersuchten Patientengutes, insbesondere hinsichtlich der peri- und postoperativen Mortalität, sowie der Auswirkungen der prophylaktischen TKR im Langzeitverlauf. Im Rahmen dieser monozentrischen, prospektiven Studie wurden 264 Patienten eingeschlossen, welche sich im Zeitraum von 2009 bis 2015 einer TKR im Rahmen einer MKR an der Klinik für Thorax- Herz- und thorakaler Gefäßchirurgie des Universitätsklinikums Frankfurt am Main unterzogen. Das Patientenkollektiv wurde nachfolgend in zwei Gruppen unterteilt, nach dem Schweregrad der präoperativ bestehenden TI in eine „prophylaktische” pTKR-Gruppe bei einer TI < 2 und eine „therapeutische” tTKR-Gruppe bei einer TI ≥ 2. Primärer Endpunkt war die Erfassung der Früh- und Spätmortalität. Sekundäres Endziel war die Untersuchung des postoperativen Verlaufes der TI, sowie der Verlauf der kardialen Funktion. Die 30-Tages-Mortalität betrug 12% in der pTKR-Gruppe und 17% in der tTKR-Gruppe. Der wichtigste Einflussfaktor war die EKZ-Dauer. Der Unterschied zwischen den beiden Gruppen war statistisch nicht signifikant. Im Vergleich zu anderen Studien zeigte sich eine höhere 30-Tages-Mortalität, jedoch handelte es sich bei diesen zumeist um einen isolierten Eingriff an der Mitralklappe, mit einer deutlich kürzeren EKZ. Hinsichtlich der Gesamtmortalität zeigte sich ein 1-, 5- und 7-Jahres-Überleben in der pTKR-Gruppe von 81%, 66% und 56%, sowie 65%, 52% und 41% in der tTKR-Gruppe. Die höhere Gesamtmortalität dieser Studie im Vergleich zu anderen Arbeiten ist durch das deutlich ältere und multimorbide Patientenkollektiv erklärt. Bei dem Vergleich von Patienten, welche im Rahmen dieser Studie eine prophylaktische TKR erhielten gegenüber den Patienten, bei welchen im Rahmen von Vergleichsarbeiten bewusst auf eine TKR verzichtet wurde (NTKR-Gruppe), zeigte sich ein verbessertes Langzeitüberleben der pTKR-Gruppe. Bei den Arbeiten, bei welchen kein Überlebensvorteil unserer pTKR-Gruppe, gegenüber deren NTKR-Gruppe gezeigt werden konnte, zeigten sich dennoch positive Effekte der begleitenden Klappenoperation. Die erhöhte Frühsterblichkeit dieser Arbeit ist dem Umstand geschuldet, dass durch die zur Indikationsstellung herangezogenen Risikofaktoren ein Hochrisikokollektiv selektioniert wurde, mit einer konsekutiv erhöhten Sterblichkeit auch abhängig vom Ausmass der TI. Bereits in der Vergangenheit konnte eine Verbesserung des Langzeitüberlebens durch die Durchführung einer TKR bei einer TI ≥ 2, gegenüber dem Verzicht auf diese nachgewiesen werden. Anhand des Vergleiches von anderen Arbeiten mit der vorliegenden Arbeit konnte dieser Überlebensvorteil auch für die Durchführung einer pTKR bei einer TI < 2 gezeigt werden. Zudem konnte dargelegt werden, dass es sich bei der begleitenden pTKR um eine effektive und dauerhafte Methode zur Vermeidung einer Klappeninsuffizienz handelt. Somit konnte gezeigt werden, dass auch bei Patienten mit einer TI < 2 eine additiven TKR die Entwicklung einer späten TI verhindert.
Knowledge is limited as to how prior SARS-CoV-2 infection influences cellular and humoral immunity after booster-vaccination with bivalent BA.4/5-adapted mRNA-vaccines, and whether vaccine-induced immunity correlates with subsequent infection. In this observational study, individuals with prior infection (n=64) showed higher vaccine-induced anti-spike IgG antibodies and neutralizing titers, but the relative increase was significantly higher in non-infected individuals (n=63). In general, both groups showed higher neutralizing activity towards the parental strain than towards Omicron subvariants BA.1, BA.2 and BA.5. In contrast, CD4 or CD8 T-cell levels towards spike from the parental strain and the Omicron subvariants, and cytokine expression profiles were similar irrespective of prior infection. Breakthrough infections occurred more frequently among previously non-infected individuals, who had significantly lower vaccine-induced spike-specific neutralizing activity and CD4 T-cell levels. Thus, the magnitude of vaccine-induced neutralizing activity and specific CD4 T-cells after bivalent vaccination may serve as a correlate for protection in previously non-infected individuals.
Graph data is an omnipresent way to represent information in machine learning. Especially, in neuroscience research, data from Diffusion-Tensor Imaging (DTI) and functional Magnetic Resonance Imaging (fMRI) is commonly represented as graphs. Exploiting the graph structure of these modalities using graph-specific machine learning applications is currently hampered by the lack of easy-to-use software. PHOTONAI Graph aims to close the gap between domain experts of machine learning, graph experts and neuroscientists. Leveraging the rapid machine learning model development features of the Python machine learning API PHOTONAI, PHOTONAI Graph enables the design, optimization, and evaluation of reliable graph machine learning models for practitioners. As such, it provides easy access to custom graph machine learning pipelines including, hyperparameter optimization and algorithm evaluation ensuring reproducibility and valid performance estimates. Integrating established algorithms such as graph neural networks, graph embeddings and graph kernels, it allows researchers without significant coding experience to build and optimize complex graph machine learning models within a few lines of code. We showcase the versatility of this toolbox by building pipelines for both resting–state fMRI and DTI data in the hope that it will increase the adoption of graph-specific machine learning algorithms in neuroscience research.
Stefan Zweig entwickelt in "Castellio gegen Calvin" (1936) die These, dass Machtformen, die Lebensprozesse mit Zwang regulieren und elementare Bedürfnisse dauerhaft unterdrücken, Widerstände hervorrufen, die letztlich die Emanzipation und Entfaltung des Unterdrückten forcieren. Am Beispiel des Liberalismus ursprünglich autoritär geprägter calvinistischer Gesellschaften veranschaulicht der 1934 nach London emigrierte Autor die Dialektik von Unterdrückung und deren Überwindung. Er thematisiert ein Verhältnis von Leben und Politik, das Michel Foucault mit dem Begriff der "Bio-Politik" bezeichnet. Dieser Begriff, der vor allem in jüngerer Vergangenheit vermehrt Aufmerksamkeit erfährt, beschreibt eine spezielle Form politischer Herrschaft, die sich durch "verschiedenste Techniken zur Unterwerfung der Körper und Kontrolle der Bevölkerungen" kennzeichnet. Die Anwendung dieser 'biopolitischen' Disziplinierungstechniken bringt nach Foucault einen speziellen Machttyp, die "Bio-Macht", hervor, die sich zu einem Prinzip aller modernen Staaten entwickelt und somit nicht nur auf Extremfälle zu beschränken ist. [...] In diesem Beitrag soll untersucht werden, inwiefern Stefan Zweig in "Castellio gegen Calvin" anhand der Darstellung des Calvin'schen Gottesstaats biopolitische Herrschaft thematisiert. Ausgehend von einigen Überlegungen zu dessen Komposition und der Schwierigkeit einer eindeutigen gattungstheoretischen Zuordnung wird Zweigs Darstellung von Calvins Herrschaft hinsichtlich der politischen Unterwerfung und Kontrolle des Lebens analysiert. Die Untersuchung schließt mit Blick auf den parabolischen Charakter des Textes, der eine Erklärung dafür bietet, warum Zweigs Darstellung des Calvin'schen Gottesstaats aus dem 16. Jahrhundert Charakteristika eines Machttyps aufweist, der sich nach Foucault erst Mitte des 18. Jahrhunderts entwickelt.
Die Biopolitik ist in den letzten Jahrzehnten zu einem grundlegenden Studienbereich für das Verständnis unserer politischen Modernität und zu einem wertvollen Instrument für die Analyse von literarischen Texten geworden. In der aktuellen Literatur zu Günter Grass' Roman "Die Blechtrommel" (1959) findet man jedoch keine ausführliche Studie, die eine biopolitische Perspektive als hermeneutische Annäherungsmethode einnimmt. Eine Analyse von "Die Blechtrommel" vor dem theoretischen Rahmen der Biopolitik ist aber für eine umfassende Lektüre des Textes notwendig, wie im Folgenden belegt wird. [...] Um sich der nationalsozialistischen Biopolitik zu widersetzen und bürgerliche Rationalitätsparadigmen zu untergraben, lässt Grass verschiedene Facetten der Populärkultur (insbesondere Volks- und Kunstmärchen), die der Nationalsozialismus entweder unterdrückte oder für seine Ideologie manipulierte, wieder aufleben. Strukturell und inhaltlich verbindet Grass' Roman die Verfolgung 'entarteter' Kunst mit der Verfolgung und Ausrottung 'entarteter' Menschen, für die der verfolgte Zwerg Oskar zur zentralen Metapher und Stimme wird. Der groteske Körper Oskars bildet in seinem Überleben und subversiven Praktiken gegen die diktatorische Autorität einen alternativen Diskurs gegen NS-Körperideale, die vor allem von Rezeptionen der klassischen Kunst beeinflusst waren. Durch eine Reihe intertextueller Bezüge zur Märchen-Tradition und seine Ästhetik des Grotesken erschafft der Roman eine irrationale Gegenkultur, die sich dem zerstörenden Rationalismus der Nazi-Biopolitik und der Kontinuität dieses Eugenik Gedankens im Nachkriegsdeutschland entgegenstellt, wobei gleichzeitig die Absurdität der Stunde Null entlarvt wird.
Das ZfL dürfte die einzige Institution gewesen sein, der er je nähergetreten ist. Als Senior Fellow schätzte er vor allem den Zugang zur Bibliothek des Hauses, wo man ihn häufig im Gespräch fand mit den Bibliothekarinnen oder anderen Besuchern, stets neugierig, mitteilungsfreudig, und meistens heiter. Manche Jüngere haben erst im Laufe der Zeit erfahren, dass der muntere ältere Herr so berühmt war. Ansonsten pflegte er gewissenhaft Abstand zu allem Institutionellen. Die Position des nicht direkt involvierten Beobachters war ihm schon während des Studiums in Frankfurt a.M. und Berlin zur zweiten Natur geworden. Sie befähigte ihn zeitlebens zu oft überraschenden, häufig provozierenden Ansichten, die sich mit Eitelkeit nicht vertrugen. Wolfgang konnte sehr charmant sein, aber er war vollständig uneitel. [...] Als Ein-Mann-Forschungsinstitution hat Wolfgang ein denkbar breites Themenfeld bearbeitet. Es reicht von der Geschichte der Eisenbahnreise (1977) über die Bedeutung der Genussmittel (1980) und die Elektrifizierung der Großstädte (1983) bis zu Analysen über die Kulturpolitik in Berlin in der kurzen Spanne nach dem Ende des Zweiten Weltkriegs und vor dem Fall des Eisernen Vorhangs (1995), die Kultur der Niederlage (2001) und das Verschwinden des geordneten Rückzugs aus der Geschichte der Kriege (2019). In Technik-, Mentalitäts-, Intellektuellen- oder Militärgeschichte und auch im wohlfeilen Label der 'Kulturgeschichte' geht keines auf - wenngleich die Frage nach Gewinnern und Verlierern der Geschichte eine Konstante im Werk darstellt.
In den aktuellen Debatten um politischen Aktivismus und institutionalisierte Wissenschaft lässt sich unschwer das alte Muster 'Elfenbeinturm' vs. Engagement erkennen, das zahlreiche Auseinandersetzungen im 20. Jahrhundert geprägt hat. Angesichts dieser langen, wechselvollen und produktiven Geschichte könnte man mit dem Thema eigentlich gelassener umgehen als der aufgeregte Ton heute nahelegt. In ihrem Beitrag zu den Osteuropawissenschaften in Zeiten des Krieges wundert sich auch Nina Weller, dass längst überwunden geglaubte Fronten sich neu formieren. Henning Trüper erinnert daran, dass moderne Wissenschaft immer von politischen Instanzen wie dem Staat abhängt. Patrick Eiden-Offe stellt anhand der Frankfurter Hölderlin-Edition, deren politische Motive einen Paradigmenwechsel in der Editionswissenschaft herbeiführten, die Verträglichkeit von Politik und Wissenschaft exemplarisch unter Beweis. Er zeigt aber auch, dass dem akademischen Erfolg des Projektes dessen politische Motive zum Opfer fielen; bei der Durchsetzung neuer Editionsprinzipien blieb der politisch-aktivistische Impuls auf der Strecke. Könnte es heute umgekehrt die Wissenschaft sein, die auf der Strecke bleibt? Und wenn es so wäre, könnte das damit zusam menhängen, dass einerseits die Politik (national und auf EU-Ebene) sowie viele Förderorganisationen eine 'transformative Wissenschaft' einfordern? Und dass andererseits immer mehr junge Menschen ihre akademische Entwicklung in den Dienst dieses oder jenes Aktivismus stellen? Haben wir es mit einer bisher unbekannten Konvergenz eines Aktivismus 'von oben' und 'von unten' zu tun?
A broad range of neuropsychiatric disorders are associated with alterations in macroscale brain circuitry and connectivity. Identifying consistent brain patterns underlying these disorders by means of structural and functional MRI has proven challenging, partly due to the vast number of tests required to examine the entire brain, which can lead to an increase in missed findings. In this study, we propose polyconnectomic score (PCS) as a metric designed to quantify the presence of disease-related brain connectivity signatures in connectomes. PCS summarizes evidence of brain patterns related to a phenotype across the entire landscape of brain connectivity into a subject-level score. We evaluated PCS across four brain disorders (autism spectrum disorder, schizophrenia, attention deficit hyperactivity disorder, and Alzheimer’s disease) and 14 studies encompassing ∼35,000 individuals. Our findings consistently show that patients exhibit significantly higher PCS compared to controls, with effect sizes that go beyond other single MRI metrics ([min, max]: Cohen’s d = [0.30, 0.87], AUC = [0.58, 0.73]). We further demonstrate that PCS serves as a valuable tool for stratifying individuals, for example within the psychosis continuum, distinguishing patients with schizophrenia from their first-degree relatives (d = 0.42, p = 4 x 10−3, FDR-corrected), and first-degree relatives from healthy controls (d = 0.34, p = 0.034, FDR-corrected). We also show that PCS is useful to uncover associations between brain connectivity patterns related to neuropsychiatric disorders and mental health, psychosocial factors, and body measurements.
In response to pathogen infection, gasdermin (GSDM) proteins form membrane pores that induce a host cell death process called pyroptosis1–3. Studies of human and mouse GSDM pores reveal the functions and architectures of 24–33 protomers assemblies4–9, but the mechanism and evolutionary origin of membrane targeting and GSDM pore formation remain unknown. Here we determine a structure of a bacterial GSDM (bGSDM) pore and define a conserved mechanism of pore assembly. Engineering a panel of bGSDMs for site-specific proteolytic activation, we demonstrate that diverse bGSDMs form distinct pore sizes that range from smaller mammalian-like assemblies to exceptionally large pores containing >50 protomers. We determine a 3.3 Å cryo-EM structure of a Vitiosangium bGSDM in an active slinky-like oligomeric conformation and analyze bGSDM pores in a native lipid environment to create an atomic-level model of a full 52-mer bGSDM pore. Combining our structural analysis with molecular dynamics simulations and cellular assays, our results support a stepwise model of GSDM pore assembly and suggest that a covalently bound palmitoyl can leave a hydrophobic sheath and insert into the membrane before formation of the membrane-spanning β-strand regions. These results reveal the diversity of GSDM pores found in nature and explain the function of an ancient post-translational modification in enabling programmed host cell death.

Background: Prostate cancer is a major health concern in aging men. Paralleling an aging society, prostate cancer prevalence increases emphasizing the need for efcient diagnostic algorithms.
Methods: Retrospectively, 106 prostate tissue samples from 48 patients (mean age,
66 ± 6.6 years) were included in the study. Patients sufered from prostate cancer (n = 38) or benign prostatic hyperplasia (n = 10) and were treated with radical prostatectomy or Holmium laser enucleation of the prostate, respectively. We constructed tissue microarrays (TMAs) comprising representative malignant (n = 38) and benign (n = 68) tissue cores. TMAs were processed to histological slides, stained, digitized and assessed for the applicability of machine learning strategies and open–source tools in diagnosis of prostate cancer. We applied the software QuPath to extract features for shape, stain intensity, and texture of TMA cores for three stainings, H&E, ERG, and PIN-4. Three machine learning algorithms, neural network (NN), support vector machines (SVM), and random forest (RF), were trained and cross-validated with 100 Monte Carlo random splits into 70% training set and 30% test set. We determined AUC values for single color channels, with and without optimization of hyperparameters by exhaustive grid search. We applied recursive feature elimination to feature sets of multiple color transforms.
Results: Mean AUC was above 0.80. PIN-4 stainings yielded higher AUC than H&E and
ERG. For PIN-4 with the color transform saturation, NN, RF, and SVM revealed AUC of 0.93 ± 0.04, 0.91 ± 0.06, and 0.92 ± 0.05, respectively. Optimization of hyperparameters improved the AUC only slightly by 0.01. For H&E, feature selection resulted in no increase of AUC but to an increase of 0.02–0.06 for ERG and PIN-4.
Conclusions: Automated pipelines may be able to discriminate with high accuracy between malignant and benign tissue. We found PIN-4 staining best suited for classifcation. Further bioinformatic analysis of larger data sets would be crucial to evaluate the reliability of automated classifcation methods for clinical practice and to evaluate potential discrimination of aggressiveness of cancer to pave the way to automatic precision medicine.
Measurements of the production of electrons from heavy-flavour hadron decays in pp collisions at s√=13 TeV at midrapidity with the ALICE detector are presented down to a transverse momentum (pT) of 0.2 GeV/c and up to pT=35 GeV/c, which is the largest momentum range probed for inclusive electron measurements in ALICE. In p−Pb collisions, the production cross section and the nuclear modification factor of electrons from heavy-flavour hadron decays are measured in the pT range 0.5<pT<26 GeV/c at sNN−−−√=8.16 TeV. The nuclear modification factor is found to be consistent with unity within the statistical and systematic uncertainties. In both collision systems, first measurements of the yields of electrons from heavy-flavour hadron decays in different multiplicity intervals normalised to the multiplicity-integrated yield (self-normalised yield) at midrapidity are reported as a function of the self-normalised charged-particle multiplicity estimated at midrapidity. The self-normalised yields in pp and p−Pb collisions grow faster than linear with the self-normalised multiplicity. A strong pT dependence is observed in pp collisions, where the yield of high-pT electrons increases faster as a function of multiplicity than the one of low-pT electrons. The measurement in p−Pb collisions shows no pT dependence within uncertainties. The self-normalised yields in pp and p−Pb collisions are compared with measurements of other heavy-flavour, light-flavour, and strange particles, and with Monte Carlo simulations.
The Heidelberg Ion-Beam Therapy Centre (HIT) provides proton, helium, and carbon-ion beams with different energies and intensities for cancer treatment and oxygen-ion beams for experiments. For several experiments and possible future applications, such as helium ion beam radiography, a low-intensity ion beam monitor integrated into the dose delivery feedback system for the accelerator control is a necessary pre-requisite. The updated 2D prototype for this purpose consists of scintillating fibres with enhanced radiation hardness, silicon photomultipliers (SiPMs) to amplify the emitted light, and a dedicated front-end readout system (FERS) to process and record the generated signals. This setup was tested successfully on monitoring ion-beam position and profile horizontally and vertically, as well as the beam intensity, for all four ion types with energies from 50 to 430 MeV/u and intensities from 1E2 to 1E7 ions/s. Additionally, time-of-arrival (ToA) measurements on single ions have been successfully performed for a limited intensity range, allowing for ion tracking in a further update. This will reduce noise, and will also improve the accuracy and usability of ion radiography.
The negative effect of fossil-based industrial processes on the environment, especially the contribution to global warming by emitting greenhouse gases such as CO2 causes a global threat to mankind. Therefore, technologies are demanded by the society for a sustainable and environmentally friendly economy. The biotechnological use of sugar-based feedstocks to produce valuable products are in conflict with, for example, food production. In order to overcome this issue, waste products such as syngas (H2, CO and CO2) or CO2 taken from the atmosphere are of increasing interest for biotechnological applications. Acetogenic bacteria are already used at industrial scale to produce sustainable and environmentally friendly biofuels from syngas. A promising candidate due to its physiological flexibility is the thermophilic acetogen Moorella thermoacetica. In contrast to most acetogens M. thermoacetica is not restricted to one energy conserving system. In addition to the Ech complex, cytochromes and quinones may be involved in energy conservation by, for example, DMSO respiration. The extra energy conserved can be used to form highly valuable but energy demanding products. In this review we give insights into the physiology of this acetogen, the current state of the art of M. thermoacetica as a platform for biotechnological applications and discuss future perspectives.
Affine Bruhat--Tits buildings are geometric spaces extracting the combinatorics of algebraic groups. The building of PGL parametrizes flags of subspaces/lattices in or, equivalently, norms on a fixed finite-dimensional vector space, up to homothety. It has first been studied by Goldman and Iwahori as a piecewise-linear analogue of symmetric spaces. The space of seminorms compactifies the space of norms and admits a natural surjective restriction map from the Berkovich analytification of projective space that factors the natural tropicalization map. Inspired by Payne's result that the analytification is the limit of all tropicalizations, we show that the space of seminorms is the limit of all tropicalized linear embeddings ι:Pr↪Pn and prove a faithful tropicalization result for compactified linear spaces. The space of seminorms is in fact the tropical linear space associated to the universal realizable valuated matroid.
Seit den 1970er Jahren haben sich die Universitäten für Senior*innen geöffnet; anfänglich nur durch die Öffnung bereits vorhandener Studiengänge, mehr und mehr aber auch durch gezielte Bildungsangebote für die Zielgruppe Älterer. Die „Universität des 3. Lebensalters“ (U3L) an der Universität Frankfurt bietet seit 40 Jahren Angebote für Ältere und hat daher Einblicke in die Bildungsbedürfnisse dieser Zielgruppe im Wandel der Zeit gesammelt: Überwog früher das Nachholen akademischer Bildung als Motiv für die Teilnahme, so geht es den sogenannten „Babyboomern“ (Jahrgänge 1955-1964) vielfach um Selbstreflexion von Lebensgestaltung und Lebensvollzug. Am Beispiel der beiden mehrjährigen Lehr-/Lern-Projekte „Biografie und Gesellschaft“ und „Männlichkeiten im Altern“ zeigt der Autor im vorliegenden Beitrag, wie eine teilnehmer*innenorientierte Seminargestaltung für diese Zielgruppe didaktisch erfolgen kann: die Inhalte in allen Phasen der didaktischen Planung entlang der biografisch begründeten Interessen der Teilnehmer*innen ausrichten, die Seminare partizipativ anlegen, eigene und fremde Erfahrungen einbeziehen, Emotionen achtsam aufgreifen und reflektieren. Als Lehrender beschreibt er sich als neugieriger und interessierter Ermöglicher und Begleiter von Lernprozessen, der sich auch selbst als alternder Mit Mensch einbringt.
Homeownership rates differ widely across European countries. We document that part of this variation is driven by differences in the fraction of adults co-residing with their parents. Comparing Germany and Italy, we show that in contrast to homeownership rates per household, homeownership rates per individual are very similar during the first part of the life cycle. To understand these patterns, we build an overlapping-generations model where individuals face uninsurable income risk and make consumption-saving and housing tenure decisions. We embed an explicit intergenerational link between children and parents to capture the three-way trade-off between owning, renting, and co-residing. Calibrating the model to Germany we explore the role of income profiles, housing policies, and the taste for independence and show that a combination of these factors goes a long way in explaining the differential life-cycle patterns of living arrangements between the two countries.
Biotechnological processes offer better production conditions for a wide variety of goods of industrial interest. The production of aromatic compounds, for example, involves molecules of great value for cosmetic, plastic, agrochemical and pharmaceutic industries. However, the yield of such processes frequently prevents a proper implementtation that would allow the replacement of traditional production processes.
Numerous rational engineering approaches have been attempted to enhance metabolic pathways associated with desired products. Unfortunately, genetic modifications and heterologous pathway expression often lead to a higher metabolic burden on the producing organisms, ultimately leading to reduced production levels and fitness.
This project utilised adaptive laboratory evolution to better understand the development of synthetic cooperative consortia, using S. cerevisiae as a model organism. Specifically, a synthetic cooperative consortium was developed around the exchange of lysine and tyrosine, which was subjected to adaptive laboratory evolution aiming to induce mutations that would improve the system’s fitness either by enhanced production or upgraded stress resistance. Consequently, the mutant strains isolated after the evolution rounds were sequenced to identify relevant variations that could be related to the growth and production phenotypes observed.
The insights derived from this project are expected to contribute to further developing synthetic cooperative consortia with utilitarian purposes.
Methods using environmental DNA to explore and analyze biodiversity from previously unexplored habitats and ecosystems have become increasingly popular in recent years. This is particularly due to the potential reduction in necessary taxonomic expertise, the opportunity to assess microorganismal communities, and decreased time investments required to cover large spatial extents. In forests, the surface of tree bark is an important habitat for epiphytic diversity. Because of the large surface area rich in micro-niches, the seasonal stability of the substrate, and the longevity of trees, tree bark surfaces provide an ideal habitat for many species. Yet, we lack a comprehensive understanding of their communities and the environmental drivers behind the community assembly. These missing links hinder the exploration of the forest microbiome as a whole and limits our understanding of functions of a large forest habitat and its connections to other forest microbiomes. With a holistic eDNA metabarcoding approach, encompassing samples of three major taxonomic groups (e.g. bacteria, fungi, and green algae), as well as simultaneous collections from multiple forest habitats we can contribute to closing these gaps and increase our knowledge of the forest microbiome.
My dissertation is set within the framework of the Biodiversity Exploratories and was conducted in four parts: I. the establishment of an eDNA metabarcoding workflow to reveal the local diversity of the bark surface microbiome; II. the upscaling of the method to large geographic and environmental gradients to uncover the drivers of the microbiome; III. the integration of soil and bark samples to investigate compositional differences in two important forest habitats; IV. the evaluation of eDNA metabarcoding as a tool for biodiversity assessments of lichen diversity in forests.
In the first part, I developed a simple, cost-effective and fast sampling strategy to acquire eDNA samples from the bark of trees in forest ecosystems. Using readily available medical-specimen-collection swabs I sampled bark surfaces of individual trees in Central German forests and used metabarcoding to amplify marker genes of green algae, fungi and bacteria. From the sequencing reads I calculated the first diversity estimates of the major organismal groups of bark surface microbiomes from Central European forests. Overall the methodology produced reliable results, allowing for an expanded sampling in the second part.
In the second part of the dissertation, I expanded the sampling based on the results of part one. I collected bark surface samples from the three regions of the Biodiversity Exploratories covering large spatial and environmental gradients representative for Central European forests. The collection included composite samples from 150 plots and over 750 trees. Utilizing measurements of climatic and forest structure variables provided by the Biodiversity Exploratories, as well as my own community data, I identified the biotic and abiotic drivers behind alpha and beta diversity of the bark surface microbiome.
In the third part, I studied the differences between the bark surface as an unexplored and the soil as an example of a well characterized forest microbiome. Using only the fungal part of the large sampling campaign and soil samples obtained from the same plots at the same time, I assessed the commonalities and differences of the micro-communities of these distinct forest niches. Furthermore, I included two coniferous and one deciduous tree species to examine, if the effect of tree species, previously shown for soil microbiomes, also holds true for the bark surface.
In the last part of my dissertation, I used eDNA in a more applied way as a tool in biodiversity assessments of lichenized fungi. I compared the results from eDNA metabarcoding to an expert floristic mapping conducted in the same plots in 2007/2008. I assigned functional guilds to the fungal taxa obtained in the large sampling campaign and used a subset that was assigned as lichenized fungi.
In conclusion, I showed that eDNA metabarcoding is a valuable tool to reveal the unknown diversity of microorganisms in forest ecosystems. In particular, my results advance our understanding of the bark surface microbiome, an underexplored habitat within forests. The tightly linked interactions of the three major microbial groups underline that studies need to take holistic approaches across multiple taxonomic groups to deepen our understanding of processes governing the assembly of microbiomes. Results from my dissertation may serve as a foundation to inform hypotheses addressing the functions of forest microbiomes. The massive diversity data collected may also contribute to closing the gap in our understanding of macro-organisms and micro-organisms with respect to diversity distributions and patterns of richness, and serve as a baseline for predictions of biodiversity responses under future anthropogenic change.
Untersuchung der Expression von Wachstumsfaktoren in reseziertem Hirngewebe von Epilepsiepatienten

Hintergrund: Die Epilepsie gehört zu den häufigsten chronischen neurologischen Erkrankungen beim Menschen. Bei Patienten mit mesialer TLE und Hippocampussklerose besteht die höchste Wahrscheinlichkeit, eine medikamentöse Therapierefraktärität zu entwickeln. Die Ursache der Hippocampussklerose sowie die ursächlichen Mechanismen sind nicht bekannt. Allerdings kann eine initiale Schädigung, wie etwa komplizierte Fieberkrämpfe im Kindesalter, Schädel-Hirn-Traumata, Schlaganfälle, entzündliche Prozesse oder Ähnliches, für die Entwicklung einer Hippocampussklerose prädisponieren. Diese kann anschließend nach einer klinisch stummen Latenzperiode zur Entwicklung spontaner epileptischer Anfälle und der Diagnose einer Epilepsie führen. Im Rahmen der Epileptogenese, also der Entstehung und Progression der Epilepsie kommt es zu Wachstumsprozessen, weshalb eine Beteiligung von neurotrophen Wachstumsfaktoren naheliegend war. Das Ziel dieser Arbeit war die vergleichende Untersuchung resezierter Hippocampi auf Wachstumsfaktoren, um semiquantitative Daten zu deren Verteilung bei Epilepsiepatienten zu erhalten. Des Weiteren war die Korrelation mit den klinischen Daten der Patienten von besonderem Interesse, da so Hinweise auf mögliche Zusammenhänge zwischen dem klinischen Erscheinungsbild und der Expression der Wachstumsfaktoren gewonnen werden konnten.
Methoden: Bei dem in der vorliegenden Arbeit untersuchten Gewebe handelt es sich um Hippocampi von 21 Patienten mit TLE, die epilepsiechirurgisch therapiert wurden. Die Schnitte der paraffinierten Hippocampi wurden mittels Immunhistochemie auf die Wachstumsfaktoren BDNF, FGF2, GDNF, GMFB und PDGF-B untersucht. Im Anschluss wurden die Schnitte gescannt und die Zellen mittels eines Algorithmus identifiziert und ausgewertet. Diese experimentellen Daten wurden anschließend mit den klinischen Daten der Patienten korreliert.
Ergebnisse: Es fand sich eine signifikante Korrelation zwischen der Expression von GMFB und dem postoperativen Outcome der Patienten. Des Weiteren fanden sich auch Korrelationen zwischen der präoperativen Anfallsfrequenz und der Expression von BDNF sowie GDNF. Auch die Epilepsiedauer korrelierte mit der Expression von BDNF. Zudem fanden sich Korrelationen zwischen den Ergebnissen der neuropsychologischen Testungen und der Expression von BDNF, sowie PDGF-B.
Diskussion: Die vorliegende Arbeit liefert einige Daten, die Hinweise für nachfolgende Untersuchungen geben können. Sowohl für die Anfallsfrequenz, als auch für die Epilepsiedauer fanden sich signifikante Korrelationen mit BDNF. Beides ist passend zu den vermuteten und zum Teil in der Literatur beschriebenen Mechanismen im Rahmen der Epilepsie, also einer postiktalen Hochregulation von Wachstumsfaktoren beziehungswiese des Zugrundegehens von Zellen im Verlauf der Erkrankung und damit zu einer reduzierten Expression von Wachstumsfaktoren. Geschlechterabhängige Unterschiede in der Expression der Wachstumsfaktoren fanden sich, passend zu der vorhandenen Literatur, nicht. Interessant ist, dass sowohl Geschlechtshormone als auch anfallssuppressive Medikamente einen Einfluss auf die Expression der Wachstumsfaktoren haben können.
Bis heute ist kein Biomarker bekannt, der eine Vorhersage über den Erfolg einer operativen Therapie bei therapierefraktären TLE treffen kann. Da meine Daten eine Korrelation von GMFB und dem postoperativen Outcome zeigen, bietet es sich für weitere Untersuchungen an, GMFB als präoperativen Biomarker zu nutzen. zu können, wäre eine einfachere Probengewinnung beispielsweise aus Blut, Liquor, Urin oder Speichel notwendig. Im Sinne einer „Liquid Biopsy“ könnte so der Erfolg einer chirurgischen Therapie weiter objektiviert werden, was die Entscheidungsfindung einfacher und risikoärmer gestalten würde.
