Im Wintersemester 2022/23 wurde die dritte universitätsweite Studierendenbefragung durchgeführt, die für Studierende ein wichtiges Instrument der Partizipation an der Qualitätsentwicklung in Studium und Lehre ist. Aus diesem Grund gebührt den 7765 Studierenden, die den umfassenden Fragebogen mit 314 Basisfragen sowie zusätzlichen fachbereichsspezifischen Fragen beantwortet haben, besonderer Dank. Die Ergebnisse der Befragung können in den Berichten auf www.studierendenbefragung.uni-frankfurt.de nachgelesen werden.
Der unter der Ägide der Freunde und Förderer der Goethe-Universität von der Paul Ehrlich-Stiftung ausgelobte Paul Ehrlich-und-Ludwig Darmstaedter-Preis ist die renommierteste Auszeichnung, die in Deutschland für medizinische Forschung verliehen wird. Den mit 120.000 Euro dotierten Preis nahm in der Frankfurter Paulskirche in diesem Jahr der Arzt und Immunologe Prof. Dennis L. Kasper (81) von der Harvard Medical School entgegen. Er hat die ersten Wörter der biochemischen Sprache entdeckt, mit der Darmbakterien unserem Immunsystem zu einer gesunden Entwicklung verhelfen. Den mit 60.000 Euro dotierten Nachwuchspreis erhielt der Chemiker Dr. Johannes Karges (31) von der Ruhr-Universität Bochum für die Entwicklung eines Verfahrens zur ferngesteuerten Tumortherapie.
Das erziehungswissenschaftliche Projekt »InterCare« will erforschen, wie junge Menschen die Doppelbelastung von Ausbildung/Studium und Pflege bewältigen. Offizieller Start des Projekts, das über vier Jahre hinweg mit 1,2 Millionen Euro von der VolkswagenStiftung gefördert wird, ist im Oktober 2024. Um den eigenen Beitrag zur sozial-ökologischen Transformation zu konkretisieren, entwickelt die Goethe-Universität in den kommenden Monaten eine Nachhaltigkeitsstrategie. Die Strategie soll klare Ziele, Maßnahmen und Verantwortlichkeiten definieren und das Bewusstsein für Nachhaltigkeit an der Universität, aber auch darüber hinaus, stärken.
Wie kommen Wissenschaft und Praxis zueinander? Unter dem Motto »Bridging the gap« stellte das Forschungsinstitut Gesellschaftlicher Zusammenhalt (FGZ) am 25. April auf einem Festival neue Formate der Wissenschaftskommunikation vor. Der UniReport sprach mit den FGZ-Referenten für Wissenstransfer Katja Maasch und Manuel Steinert.
Lifestyle factors—such as diet, physical activity (PA), smoking, and alcohol consumption—have a significant impact on mortality as well as healthcare costs. Moreover, they play a crucial role in the development of type 2 diabetes mellitus (DM2). There also seems to be a link between lifestyle behaviours and insulin resistance, which is often a precursor of DM2. This study uses an enhanced Healthy Living Index (HLI) integrating accelerometric data and an Ecological Momentary Assessment (EMA) to explore differences in lifestyle between insulin-sensitive (IS) and insulin-resistant (IR) individuals. Moreover, it explores the association between lifestyle behaviours and inflammation. Analysing data from 99 participants of the mPRIME study (57 women and 42 men; mean age 49.8 years), we calculated HLI scores—ranging from 0 to 4— based on adherence to specific low-risk lifestyle behaviours, including non-smoking, adhering to a healthy diet, maximally moderate alcohol consumption, and meeting World Health Organization (WHO) PA guidelines. Insulin sensitivity was assessed using a Homeostatic Model Assessment of Insulin Resistance (HOMA-IR) and C-reactive protein (CRP) levels were used as a proxy for inflammation. Lifestyle behaviours, represented by HLI scores, were significantly different between IS and IR individuals (U = 1529.0; p = 0.023). The difference in the HLI score between IR and IS individuals was mainly driven by lower adherence to PA recommendations in the IR group. Moreover, reduced PA was linked to increased CRP levels in the IR group (r = −0.368, p = 0.014). Our findings suggest that enhancing PA, especially among individuals with impaired insulin resistance, holds significant promise as a preventive strategy.
Scalibregmatidae Malmgren, 1867 is a family of annelids, with only few taxa reported from Brazil. In this work, we describe two new species of Asclerocheilus Ashworth, 1901. The new species A. geiseae sp. nov. and A. blakei sp. nov. were collected during scientific expeditions to the Espírito Santo and Campos sedementary basins. These expeditions were coordinated by PETROBRAS/CENPES. The species A. geiseae sp. nov. is unique among its congeners by the presence of acicular spines with rounded and curved tips in two rows. The second species, A. blakei sp. nov., differs from its congeners by the combination of the presence of a trapezoidal prostomium, acicular spines on chaetigers 1–3, lyrate chaetae from chaetiger 3 on notopodia and 2 on neuropodia. Moreover, we report the species A. tropicus Blake, 1981 from the localities sampled on this study. We also provide an identification key to all species of Asclerocheilus of the world.
Five new species of the spider genus Heteropoda Latreille, 1804 (Araneae: Sparassidae) from China

Five new species of Heteropoda (Araneae, Sparassidae, Heteropodinae) are described from China: H. bawanglingensis sp. nov. (female; Hainan), H. dulongensis sp. nov. (male, female; Yunnan), H. hainanensis sp. nov. (male, female; Hainan), H. longa sp. nov. (female; Guizhou), and H. vaginalis sp. nov. (female; Yunnan). We provide descriptions and illustrations for each species as well as a distribution map in the current paper.
A taxonomic study on the Afrotropical genera Afrepipona Giordani Soika, 1965 and Afrepsilon gen. nov. is presented. Twenty new species are described, of which 15 are in Afrepipona (Afrepipona anomala sp. nov., Afrepipona cellularis sp. nov., Afrepipona clonata sp. nov., Afrepipona cuprea sp. nov., Afrepipona lamellata sp. nov., Afrepipona lamptula sp. nov., Afrepipona lobulata sp. nov., Afrepipona meridionalis sp. nov., Afrepipona occidentalis sp. nov., Afrepipona orientalis sp. nov., Afrepipona punctatissima sp. nov., Afrepipona scabra sp. nov., Afrepipona segregata sp. nov., Afrepipona ulterior sp. nov., Afrepipona vulcanica sp. nov.) and five are in Afrepsilon (Afrepsilon aterrimum gen. et sp. nov., Afrepsilon ferrugineoaureum gen. et sp. nov., Afrepsilon hybridum gen. et sp. nov., Afrepsilon minor gen. et sp. nov., and Afrepsilon pictum gen. et sp. nov.). Afrepipona lamptoensis Giordani Soika, 1965 is withdrawn from synonymy with A. angusta (de Saussure, 1863) and subsequently revalidated. Afrepipona unifasciata Gusenleitner, 2012 is transferred to the genus Antodynerus de Saussure, 1855 (Antodynerus unifasciatus comb. nov.). Keys to all known species are provided.
Description of four new species of the Neotropical genus Scione Walker, 1850 (Diptera: Tabanidae)

After 94 years after the last revision of the genus and after 22 years since the last species was named, we describe four new species of Scione from Ecuador and Peru: S. cooperi sp. nov., S. gertrudeae sp. nov., S. kroeberi sp. nov. and S. merianae sp. nov. We provide diagnoses, descriptions, photographs and discussions for each of the species.
Sipunculans are a poorly studied group in the Tropical Eastern Pacific. For the Southern Mexican Pacific (SMP) there is only one record of a sipunculan species. The main objective of this work was to determine the species composition of the phylum Sipuncula present in the SMP. The study area covered three Mexican states: Guerrero, Oaxaca and Chiapas; specimens from 28 localities were examined from both intertidal and subtidal zones. A total of 551 specimens were reviewed, from which 11 species were identified. Five of them have previously been recorded in the Tropical Eastern Pacific (TEP): Apionsoma (A.) hespera comb. nov., A. (Edmondsius) pectinatum, Aspidosiphon (A.) elegans, Phascolosoma (P.) puntarenae and Themiste (T.) hennahi; four species are similar to nominal species: Sipunculus (S.) cf. polymyotus, Siphonosoma cf. vastum, Siphonosoma cf. cumanense and Phascolosoma (P.) cf. perlucens; and two new species are described: Aspidosiphon (Paraspidosiphon) cutleri sp. nov. and Aspidosiphon (Paraspidosiphon) pastori sp. nov. A checklist and an identification key for all sipunculan species from the TEP are presented. The checklist includes 53 taxa, 25 of which are questionable records. This work generated 11 new records of sipunculans in the SMP and five new records in the TEP.
Species lists play an important role in biology and practical domains like conservation, legislation, biosecurity and trade regulation. However, their effective use by non-specialist scientific and societal users is sometimes hindered by disagreements between competing lists. While it is well-known that such disagreements exist, it remains unclear how prevalent they are, what their nature is, and what causes them. In this study, we argue that these questions should be investigated using methods based on taxon concept rather than methods based on Linnaean names, and use such a concept-based method to quantify disagreement about bird classification and investigate its relation to research effort. We found that there was disagreement about 38% of all groups of birds recognized as a species, more than three times as much as indicated by previous measures. Disagreement about the delimitation of bird groups was the most common kind of conflict, outnumbering disagreement about nomenclature and disagreement about rank. While high levels of conflict about rank were associated with lower levels of research effort, this was not the case for conflict about the delimitation of bird groups. This suggests that taxonomic disagreement cannot be resolved simply by increasing research effort.

MicroRNAs (miRNAs) are critical post-transcriptional regulators in many biological processes. They act by guiding RNA-induced silencing complexes to miRNA response elements (MREs) in target mRNAs, inducing translational inhibition and/or mRNA degradation. Functional MREs are expected to predominantly occur in the 3’ untranslated region and involve perfect base-pairing of the miRNA seed. Here, we generate a high-resolution map of miR-181a/b-1 (miR-181) MREs to define the targeting rules of miR-181 in developing murine T-cells. By combining a multi-omics approach with computational high-resolution analyses, we uncover novel miR-181 targets and demonstrate that miR-181 acts predominantly through RNA destabilization. Importantly, we discover an alternative seed match and identify a distinct set of targets with repeat elements in the coding sequence which are targeted by miR-181 and mediate translational inhibition. In conclusion, deep profiling of MREs in primary cells is critical to expand physiologically relevant targetomes and establish context-dependent miRNA targeting rules.

Here, we introduce racoon_clip, a sustainable and fully automated pipeline for the complete processing of iCLIP and eCLIP data to extract RNA binding signal at single-nucleotide resolution. racoon_clip is easy to install and execute, with multiple pre-settings and fully customizable parameters, and outputs a conclusive summary report with visualizations and statistics for all analysis steps.
We perform the first investigation of the process e+e−→K+K−ψ(2S) and report its Born cross sections over a range of center-of-mass energies from 4.699 to 4.951~GeV. The measurements are carried out using several partial reconstruction techniques using data samples collected by the BESIII detector with a total integrated luminosity of 2.5~fb−1. We search for new tetraquark candidates Z±cs in the decays Z±cs→K±ψ(2S). No significant Z±cs signals are observed.
Discoveries of new species often depend on one or a few specimens, leading to delays as researchers wait for additional context, sometimes for decades. There is currently little professional incentive for a single expert to publish a stand-alone species description. Additionally, while many journals accept taxonomic descriptions, even specialist journals expect insights beyond the descriptive work itself. The combination of these factors exacerbates the issue that only a small fraction of marine species are known and new discoveries are described at a slow pace, while they face increasing threats from accelerating global change. To tackle this challenge, this first compilation of Ocean Species Discoveries (OSD) presents a new collaborative framework to accelerate the description and naming of marine invertebrate taxa that can be extended across all phyla. Through a mode of publication that can be speedy, taxonomy-focused and generate higher citation rates, OSD aims to create an attractive home for single species descriptions. This Senckenberg Ocean Species Alliance (SOSA) approach emphasises thorough, but compact species descriptions and diagnoses, with supporting illustrations and with molecular data when available. Even basic species descriptions carry key data for distributions and ecological interactions (e.g., host-parasite relationships) besides universally valid species names; these are essential for downstream uses, such as conservation assessments and communicating biodiversity to the broader public.This paper presents thirteen marine invertebrate taxa, comprising one new genus, eleven new species and one re-description and reinstatement, covering wide taxonomic, geographic, bathymetric and ecological ranges. The taxa addressed herein span three phyla (Mollusca, Arthropoda, Echinodermata), five classes, eight orders and twelve families. Apart from the new genus, an updated generic diagnosis is provided for four other genera. The newly-described species of the phylum Mollusca are Placiphorella methanophila Vončina, sp. nov. (Polyplacophora, Mopaliidae), Lepetodrilus marianae Chen, Watanabe & Tsuda, sp. nov. (Gastropoda, Lepetodrilidae), Shinkailepas gigas Chen, Watanabe & Tsuda, sp. nov. (Gastropoda, Phenacolepadidae) and Lyonsiella illaesa Machado & Sigwart, sp. nov. (Bivalvia, Lyonsiellidae). The new taxa of the phylum Arthropoda are all members of the subphylum Crustacea: Lepechinella naces Lörz & Engel, sp. nov. (Amphipoda, Lepechinellidae), Cuniculomaera grata Tandberg & Jażdżewska, gen. et sp. nov. (Amphipoda, Maeridae), Pseudionella pumulaensis Williams & Landschoff, sp. nov. (Isopoda, Bopyridae), Mastigoniscus minimus Wenz, Knauber & Riehl, sp. nov. (Isopoda, Haploniscidae), Macrostylis papandreas Jonannsen, Riehl & Brandt, sp. nov. (Isopoda, Macrostylidae), Austroniscus indobathyasellus Kaiser, Kniesz & Kihara, sp. nov. (Isopoda, Nannoniscidae) and Apseudopsis daria Esquete & Tato, sp. nov. (Tanaidacea, Apseudidae). In the phylum Echinodermata, the reinstated species is Psychropotes buglossa E. Perrier, 1886 (Holothuroidea, Psychropotidae).The study areas span the North and Central Atlantic Ocean, the Indian Ocean and the North, East and West Pacific Ocean and depths from 5.2 m to 7081 m. Specimens of eleven free-living and one parasite species were collected from habitats ranging from an estuary to deep-sea trenches. The species were illustrated with photographs, line drawings, micro-computed tomography, confocal laser scanning microscopy and scanning electron microscopy images. Molecular data are included for nine species and four species include a molecular diagnosis in addition to their morphological diagnosis.The five new geographic and bathymetric distribution records comprise Lepechinella naces Lörz & Engel, sp. nov., Cuniculomaera grata Tandberg & Jażdżewska, sp. nov., Pseudionella pumulaensis Williams & Landschoff, sp. nov., Austroniscus indobathyasellus Kaiser, Kniesz & Kihara, sp. nov. and Psychropotes buglossa E. Perrier, 1886, with the novelty spanning from the species to the family level. The new parasite record is Pseudionella pumulaensis Williams & Landschoff, sp. nov., found in association with the hermit crab Pagurus fraserorum Landschoff & Komai, 2018.
Observation of χcJ → 3(K⁺K⁻)

By analyzing (27.12±0.14)×108 𝜓(3686) events collected with the BESIII detector operating at the BEPCII collider, the decay processes 𝜒𝑐𝐽→3(𝐾+𝐾−) (𝐽=0, 1, 2) are observed for the first time with statistical significances of 8.2𝜎, 8.1𝜎, and 12.4𝜎, respectively. The product branching fractions of 𝜓(3686)→𝛾𝜒𝑐𝐽, 𝜒𝑐𝐽→3(𝐾+𝐾−) are presented and the branching fractions of 𝜒𝑐𝐽→3(𝐾+𝐾−) decays are determined to be ℬ𝜒𝑐0→3(𝐾+𝐾−)=(10.7±1.8±1.1)×10−6, ℬ𝜒𝑐1→3(𝐾+𝐾−)=(4.2±0.9±0.5)×10−6, and ℬ𝜒𝑐2→3(𝐾+𝐾−)=(7.2±1.1±0.8)×10−6, where the first uncertainties are statistical and the second are systematic.
Using (2.712±0.014)×109 ψ(3686) events collected with the BESIII detector operating at the BEPCII, we find an evidence of the ηc(2S)→K+K−η′ decay with a statistical significance of 3.1σ. Its decay branching fraction is measured to be (12.24±4.60(stat.)±2.37(syst.)±4.68(extr.))×10−4, where the first uncertainty is statistical, the second is systematic, and the third uncertainty is from the branching fraction of the ψ(3686)→γηc(2S) decay. The upper limit on the product branching fraction B[ψ(3686)→γηc(2S)]× B[ηc(2S)→K+K−η′] is set to be 1.14×10−6 at 90% confidence level. In addition, the branching fractions of χc1→K+K−η′ and χc2→K+K−η′ are updated to be (8.47±0.09(stat.)±0.47(syst.))×10−4 and (1.53±0.04(stat.)±0.08(syst.))×10−4, respectively. The precision is improved by twofold.
In the deep sea, interactions between benthic fauna and seafloor sediment primarily occur through bioturbation that can be preserved as traces (i.e. lebensspuren). Lebensspuren are common features of deep-sea landscapes and are more abundant than the organisms that produce them (i.e. tracemakers), rendering lebensspuren promising proxies for inferring biodiversity. The density and diversity relationships between lebensspuren and benthic fauna remain unclear, and contradicting correlations have been proposed (i.e. negative, positive, or even null correlations). To approach these variable correlations, lebensspuren and benthic fauna were characterized taxonomically at eight deep-sea stations in the Kuril-Kamchatka Trench area, together with two novel categories: tracemakers (specific epibenthic fauna that produce these traces) and degrading fauna (benthic fauna that can erase lebensspuren). No general correlation (overall study area) was observed between diversities of lebensspuren, tracemakers, degrading fauna, and fauna. However, a diversity correlation was observed at specific stations, showing both negative and positive correlations depending on: (1) the number of unknown tracemakers (especially significant for dwelling lebensspuren); (2) the lebensspuren with multiple origins; and (3) tracemakers that can produce different lebensspuren. Lebensspuren and faunal density were not correlated. However, lebensspuren density was either positively or negatively correlated with tracemaker densities, depending on the lebensspuren morphotypes. A positive correlation was observed for resting lebensspuren (e.g. ophiuroid impressions, Actiniaria circular impressions), while negative correlations were observed for locomotion-feeding lebensspuren (e.g. echinoid trails). In conclusion, lebensspuren diversity may be a good proxy for tracemaker biodiversity when the lebensspuren–tracemaker relationship can be reliable characterized. Lebensspuren–density correlations vary depending on the specific lebensspuren residence time, tracemaker density, and associated behaviour (rate of movement). Overall, we suggest that lebensspuren density and diversity correlations should be studied with tracemakers rather than with general benthic fauna. On a global scale, abiotic (e.g. hydrodynamics, substrate consistency) and other biotic factors (e.g. microbial degradation) may also play an important role.
This paper examines the dynamic relationship between firm leverage and risktaking. We embed the traditional agency problem of asset substitution within a multi-period model, revealing a U-shaped relationship between leverage and risktaking, evident in data from both the U.S. and Europe. Firms with medium leverage avoid risk to preserve the option of issuing safe debt in the future. This option is valuable because safe debt does not incur the expected cost of bankruptcy, anticipated by debt-holders due to future risk-taking incentives. Our model offers new insights on the interaction between companies' debt financing and their risk profiles.
Cross-predictability denotes the fact that some assets can predict other assets' returns. I propose a novel performance-based measure that disentangles the economic value of cross-predictability into two components: the predictive power of one asset's signal for other assets' returns (cross-predictive signals) and the amount of an asset's return explained by other assets' signals (cross-predicted returns). Empirically, the latter component dominates the former in the overall cross-prediction effects. In the crosssection, cross-predictability gravitates towards small firms that are strongly mispriced and difficult to arbitrage, while it becomes more difficult to cross-predict returns when market capitalization and book-to-market ratio rise.
In this study, we describe two new species of Mesobiotus based on morphological data collected through light and scanning electron microscopy. Descriptions include DNA sequences of four commonly used molecular markers (18S rDNA, 28S rDNA, ITS-2, and COI). Mesobiotus efa sp. nov. was discovered in North-West Russia and belongs to the group of species with smooth cuticle, harmsworthi-type OCA, typical Mesobiotus claws IV with unindented lunules, and egg chorion with reticulated processes in form of ‘sharp wide cones’ or ‘cones with long slender endings’, egg process bases with well-developed crone of dark thickenings without finger-like projections, and egg shell surface between the processes with ridges without reticulation, areolation or semi-areolation. It can be distinguished from all know species of this group by a unique combination of morphological and morphometric characters. Mesobiotus vulpinus sp. nov. was found in the Russian Far East, and is similar to Mesobiotus mauccii by having an egg chorion with polygonal relief. The new species can be distinguished from M. mauccii by having a narrower buccal tube, by details of oral cavity armature, and by longer egg chorion processes. Furthermore, we provide results of the phylogenetic analyses of the genus Mesobiotus conducted in this study.
Ophioderma teres (Lyman, 1860), an ophiuroid previously believed to have a wide distribution in the eastern Pacific, has been found to have an unclear taxonomic identity. While considered a well-known species, recent studies have revealed that O. teres lacks a holotype and has vague boundaries with its congeners Ophioderma teres unicolor H.L. Clark, 1940 and Ophioderma sodipallaresi Caso, 1986, as well as with two additional new morphotypes detected in Mexico and Nicaragua, causing continuous misidentifications. This study utilized an integrative taxonomy approach based on morphologic, morphometric, and molecular evidence to clarify the taxonomic status of O. teres, O. sodipallaresi, O. teres unicolor, and the two new morphotypes. Data integration led to the following results: 1) the neotype designation and redescription of O. teres; 2) the proposal of O. sodipallaresi as a junior synonym of O. teres; 3) the status change of O. unicolor stat. nov. from subspecies to species, and 4) the description of the morphotypes as the new species Ophioderma aija sp. nov. and Ophioderma bichi sp. nov. An identification key to the eastern Pacific species of Ophioderma was also developed. This work contributes to the knowledge of Ophioderma in the region, increasing the number of described species and providing resources for their accurate identification.
The monophyly of Theraphosinae is supported by both morphological and molecular phylogenies. However, intergeneric relationships often show polytomies and branches with low support. A previous phylogenetic study proposed an intergeneric relationship for the subfamily based on molecular data and divided it into three tribes: Grammostolini, Hapalopini and Theraphosini. However, not all genera of Theraphosinae were sampled, and some relationships were inferred based on morphological similarities. Regarding the Hapalopini from Brazil, the relationships of some genera are still uncertain, such as Kochiana, Catanduba, and Munduruku. In this paper, we describe four new species of Hapalopini from Brazil: Cyriocosmus paresi sp. nov., Hapalopus akroa sp. nov., H. guidonae sp. nov., and K. fukushimae sp. nov. In addition, we propose an updated diagnosis for these three genera and for the species K. brunnipes and M. bicoloratum.

In den letzten Jahren haben verschiedene pandemische Viren zu beträchtlichen Krankheits- und Todesfällen geführt. Um dieser ständigen Bedrohung entgegenzuwirken, ist es wichtig diagnostische Testsysteme und Therapien anzupassen oder neu zu etablieren. Diese Arbeit konzentriert sich auf die pandemischen Viren SARS-CoV-2 und HIV.
Der Zelleintritt von SARS-CoV-2 wird durch das Spike-Protein (S) ausgelöst, welches die Fusion der Virushülle mit der zellulären Membran bewirkt. Erste Studien haben gezeigt, dass das S-Protein eine hohe Fusionsaktivität aufweist. Aus diesem Grund sollten in dieser Arbeit neue Fusionstests etabliert werden, um potenzielle Inhibitoren der Zellfusion zu evaluieren. Im ersten Teil dieser Thesis wird die Etablierung von quantitativen Tests zur Evaluierung der Zell-Zell und Partikel-Zell Fusionsaktivität, welche durch S bewirkt wird, demonstriert.
Trotz jahrelanger Forschung können HIV-Patienten nicht geheilt werden und Virusinfektionen treten weiterhin weltweit auf. Das größte Problem bei der Entwicklung eines Heilmittels ist die frühe Bildung von Reservoirzellen während einer Infektion. Um diese Reservoirzellen zu identifizieren, wurde der Oberflächenmarker CD32a vorgeschlagen. Die Nutzung von Cas9-Nukleasen zur Inaktivierung von HIV ist in vitro erfolgreich, aber der effiziente Transfer in Reservoirzellen bleibt weiterhin herausfordernd. Im zweiten Teil dieser Thesis werden Rezeptor-gerichtete Adeno-assoziierte Vektoren (AAVs) für die HIV-Gentherapie präsentiert, die CD4 und CD32a für den Zelleintritt nutzen.
Zur Charakterisierung der Fusionsaktivität von SARS-CoV-2 wurden drei quantitative Fusionstests etabliert, welche Partikel- und Zell-Zell Fusionen berücksichtigen. Für den Partikel-Zell Fusionstest wurden lentivirale Vektoren (LV) verwendet, welche das S-Protein auf ihrer Oberfläche präsentierten. Die Transduktionseffizienz von S-LV erreichte auf Zellen, die den SARS-CoV-2 Rezeptor ACE2 exprimieren, ein Signal-zu-Hintergrund Verhältnis von über 2000. Durch die Präsentation von S auf leeren LV-Partikeln konnte die Fusion von benachbarten Zellen detektiert und quantifiziert werden („fusion-from-without“ (FFWO)). Für die Quantifizierung wurde ein Reporter-Komplementationstest etabliert. Hierbei wurden die Alpha- und Omega-Fragmente der β-Galaktosidase getrennt in zwei Zielzellpopulationen exprimiert, die beide ACE2 exprimierten. Durch die Zugabe von S-Partikeln kam es zur Fusion der Zielzellen und zur Komplementation der Alpha- und Omega-Fragmente. Die resultierende β-Galaktosidase-Aktivität konnte anschließend quantifiziert werden. Unter optimalen Versuchsbedingungen erreichte dieser Assay ein Signal-zu-Hintergrund Verhältnis von 2,7 Größenordnungen. Anschließend wurde der Komplementationstest für die Messung der Zell-Zell Fusion verwendet. In diesem Test exprimierten Effektorzellen S und das Alpha-Fragment, Zielzellen ACE2 und das Omega-Fragment. Obwohl die S-Expression auf den Effektorzellen sehr gering war, konnte dennoch eine signifikante Fusion nachgewiesen werden. Auch hier konnte unter optimalen Versuchsbedingungen ein hohes Signal-zu-Hintergrund Verhältnis von 2,9 Größenordnungen festgestellt werden. Nach der Etablierung der Testsysteme wurden S-spezifische Inhibitoren verwendet. Im Gegensatz zu Partikel-Zell-Fusionen wurde die Fusionsaktivität von S auf Zellen nur mäßig inhibiert. Dies deutet daraufhin, dass das Eindringen von Partikeln in Zellen wirksamer verhindert werden kann als die Ausbreitung durch Zell-Zell Fusionen.
Um AAVs spezifisch an HIV-Reservoirzellen zu binden, wurden CD4- und CD32a-spezifische DARPins („designed ankyrin repeat proteins“) in Rezeptor-verblindete AAVs eingebaut. Ebenso wurden beide DARPins gleichzeitig auf dem Kapsid präsentiert, um eine höhere Spezifität für doppelt-positive Zellen zu erreichen. Wenn diese Partikel einer Zellmischung aus CD4-, CD32a- und CD4/CD32a-exprimierenden Zellen zugesetzt wurden, transduzierten die bispezifischen Vektoren vorzugsweise doppelt-positive Zellen. Diese Präferenz war am höchsten in Zellkulturen, die stark unterrepräsentierte CD4/CD32a-exprimierende Zellen enthielten. Unter diesen Voraussetzungen erreichten bispezifische Vektoren eine bis zu 66-fach höhere Transduktionseffizienz auf CD4/CD32a-positive Zellen im Vergleich zu CD32a-exprimierenden Zellen. Darüber hinaus zeigten bispezifische AAV eine präferentielle Bindung und Transduktion von isolierten Primärzellen und Zellen in Vollblut. Selbst nach systemischer Injektion in humanisierte Mäuse wurden doppelt-positive Zellen effizienter von bispezifischen als von monospezifischen AAVs transduziert. Schließlich zeigten die generierten Vektoren, welche die Cas9 Nuklease transferierten, eine effiziente Inhibition der HIV-Replikation.
Die E3-Ubiquitinligase TRIM25 ist in verschiedenen humanen Tumoren verstärkt exprimiert, was häufig mit einer schlechten Prognose der betroffenen Patienten sowie dem Auftreten von Therapieresistenzen korreliert. Unsere Arbeitsgruppe konnte TRIM25 zuvor als Caspase-2 und -7 mRNA-bindendes Protein und negativen Regulator beider Caspasen in humanen Kolonkarzinomzellen identifizieren. Ein transienter TRIM25 Knockdown führt in Abhängigkeit der erhöhten Expression der jeweiligen Caspasen zur Sensitivierung der Kolonkarzinomzellen gegenüber Chemotherapeutika-induzierter Apoptose. Ein Ziel dieser Arbeit war, die Übertragbarkeit dieser Erkenntnisse auf einen loss-of-function-Ansatz mit stabilen TRIM25 Knockdown Zellen zu überprüfen. Die stabilen Knockdown Zellen sollten der späteren Etablierung eines Xenograftmodells dienen. Da zahlreiche TRIM Proteine bekannterweise eine vielseitige Rolle bei der Regulation sowohl onkogener als auch tumorsuppressiver Prozesse einnehmen, wurde als weitere maßgebliche Fragestellung dieser Arbeit der Einfluss von TRIM25 auf wichtige tumorigene Eigenschaften wie Proliferation, Migration, Zellzyklus und Inflammation untersucht. TRIM25-abhängige Effekte auf das Migrationsver halten von RKO-Zellen wurden im in vitro-Wundheilungsassay mit stabilen TRIM25 Knock down Zellen analysiert. Aufgrund hoher interexperimenteller Unterschiede im Migrations verhalten derselben Zellklone konnte hinsichtlich einer TRIM25-abhängigen Regulation der Migration jedoch keine eindeutige Aussage getroffen werden. Dagegen belegten die gezeigten Proliferationsassays eine signifikant vermehrte Proliferation von RKO-Zellen nach stabilem TRIM25 Knockdown. Dies legt eine tumorsuppressive Rolle von TRIM25 nahe. Durch flusszytometrische Analysen stabiler TRIM25 Knockdown und Kontrollzellen zeigten hinge gen keinen konsistenten Einfluss von TRIM25 auf den Zellzyklus von RKO-Zellen. Eine Sensitivierung von Kolonkarzinomzellen gegenüber Chemotherapeutika-induzierter Apoptose konnte auch in stabilen TRIM25 Knockdown Zellen nachgewiesen werden, während die für transiente Ansätze bekannte, TRIM25 Knockdown-abhängige Hochregulation von Caspase 2 und -7 dagegen deutlich geringer ausgeprägt war. Dies lässt vermuten, dass die Tumorzellen einer Hochregulation Zelltod-induzierender Proteine bei einem konstitutiven TRIM25Knockdown gegenregulieren. Aufgrund der aber nach wie vor nachweisbaren Sensitivierung der TRIM25 Knockdown Zellen gegenüber Apoptose, können zusätzliche, bisher noch nicht bekannte Mechanismen postuliert werden, welche zur Sensitivierung dieser Zellen gegenüber Apoptose beitragen. Die daraus abgeleitete, TRIM25-abhängige Apoptosehemmung spricht für einen „Überlebensmechanismus“, welcher maßgeblich zur Chemotherapieresis tenz von Kolonkarzinomzellen beitragen kann. Hinsichtlich eines Einflusses von TRIM25 auf entzündliche Prozesse wurden RKO-Zellen mit klassischen Aktivatoren des NLRP3-Inflammasoms stimuliert und ausgewählte Marker mittels Western Blot-Analysen nachgewiesen. TRIM25 Knockdown-abhängig war eine verminderte Spaltung der Apoptosemarker Caspase-3, -7 und PARP-1 nachweisbar. Caspase-7 und PARP-1 spielen bekanntermaßen auch im Rahmen Inflammasom-induzierter Signalprozesse eine wichtige Rolle, während die Spaltung von Caspase-3 durch das NLRP3-Inflammasom induziert werden kann und v.a. für apoptotische oder pyroptotische Prozesse verantwortlich gemacht wird. Daher kann postuliert werden, dass der stabile TRIM25 Knockdown zur Hemmung von Inflammasom-induzierten Signalprozessen führt und darüber Kolonkarzinomzellen vor Apoptose/Pyroptose geschützt werden. Umgekehrt deutet dies auf eine Aktivierung von Inflammasom-vermittelten Signalprozessen durch TRIM25 hin.
Aufgrund der hohen Inzidenz und Mortalität, der schlechten Prognose und der Entwicklung von Therapieresistenzen besteht ein dringender Bedarf in der Entwicklung neuer, verbesserter Therapien des kolorektalen Karzinoms sowie der Resensibilisierung der Krebszellen gegenüber gängigen, bereits bestehenden Therapieoptionen. Im Rahmen dieser Arbeit konnte ein onkogener Einfluss von TRIM25 auf Kolonkarzinomzellen durch Hemmung Chemotherapeutika-induzierter Apoptose nachgewiesen werden, während eine mögliche tumorigene Rolle von TRIM25 durch zusätzliche Aktivierung Inflammasom-induzierter Signalprozesse weiterer Untersuchungen bedarf. Zusammenfassend kann TRIM25 als vielversprechender therapeutischer Angriffspunkt für die Entwicklung von sowohl neuen antientzündlichen als auch tumorsuppressiven Therapien betrachtet werden.
How can older adults participate equally in digitisation processes across Europe, and what inclusive research strategies are needed? This Zine summarizes findings from a “Research Innovation Lab on Ageing in a Digital Age”, funded by the VolkswagenStiftung, aiming to bring together 29 docs and postdocs anchored in 26 different disciplines coming from 11 countries, at all stages of their work, to address cutting edge questions relating to ageing in a digital age. Five groups worked together over five days in Frankfurt, Germany, in July 2023 in a creative and interactive hackathon, specific to developing non-technical solutions to social issues of this topic. Moreover, four distinguished experts presented keynote speeches and proposals from various conceptual, methodological and empirical perspectives.
Finanzielle Armut prägt Mobilitätspraktiken und kann dabei zum Prozess von mobilitätsbezogener sozialer Exklusion beitragen. Zu den Personen, deren Armutsrisiko besonders hoch ist, zählen in Deutschland Haushalte mit Kindern, insbesondere Alleinerziehende. Ältere Menschen haben nicht die höchste Armutsgefährdung, jedoch besteht bei ihnen das Risiko von Verharrung in Armut, da die Möglichkeiten, die finanzielle Situation aus eigener Kraft zu ändern mit zunehmendem Alter sinken.
Um ein tieferes Verständnis davon zu erhalten, wie finanzielle Armut die Mobilitätspraktiken und soziale Teilhabe von Haushalten mit Kindern sowie älteren Menschen prägt, wurden mit diesen beiden Personengruppen problemzentrierte Interviews in Ronnenberg (Region Hannover) geführt und analysiert. Die Ergebnisse belegen, dass, wenngleich alle Befragten mit ähnlich geringen finanziellen Ressourcen haushalten und Verzicht sowie Abwägungsprozesse notwendig sind, sich ihre Mobilitätspraktiken und Alltagsbewältigungsstrategien unterscheiden, was sich in zwei Typologien widerspiegelt. Erstens, eine Typologie der Mobilitätspraktiken von Haushalten mit Kindern: (i) autozentriert, (ii) autoreduziert, (iii) ÖPNV-orientiert und (iv) nichtmotorisiert. Zweitens, eine Typologie älterer Menschen anhand ihrer Mobilitätspraktiken: (i) aktive ältere Menschen mit vielseitigen sozialen Interaktionen, (ii) nachbarschaftsorientierte ältere Menschen mit lokalen Kontakten und (iii) ältere Menschen, die überwiegend zu Hause sind und wenig soziale Kontakte haben.
Um herauszufinden, inwiefern mobilitätsbezogene Barrieren der sozialen Teilhabe reduziert werden können, wurden fünf Maßnahmen bezüglich ihrer Wirkung auf die Mobilitätspraktiken einkommensarmer Haushalte mit Kindern untersucht: einerseits die Wirkung des 9-Euro-Tickets anhand von problemzentrierten Interviews mit einkommensarmen Haushalten mit Kindern, andererseits anhand von Expert:inneninterviews die Wirkung von Radlernkursen für Frauen mit Migrationshintergrund, eines Mietertickets, eines Quartierstickets und der Verbesserung der Nahraum- und Aufenthaltsqualität am Beispiel von Tempo 30. Die Ergebnisse zum 9-Euro-Ticket belegen, dass ein erschwingliches ÖPNV-Ticket erheblich zur Reduzierung mobilitätsbezogener Barrieren der sozialen Teilhabe im Armutskontext beiträgt. Die Expert:inneninterviews zeigen auf, dass eine Förderung des Umweltverbunds zielführend ist, um zu einer sozial-ökologischen Verkehrswende beizutragen und insbesondere Maßnahmenbündel Wirkung auf die Reduzierung von mobilitätsbezogenen Barrieren der sozialen Teilhabe entfalten.
Die Erkenntnisse dieser Dissertation ergänzen den wissenschaftlichen Forschungstand um ein tiefergehendes Verständnis der Wirkung von finanzieller Armut auf die Mobilitätspraktiken und soziale Teilhabe von Haushalten mit Kindern und älteren Menschen und helfen dabei, Maßnahmen zur Reduzierung mobilitätsbezogener Barrieren der sozialen Teilhabe zu konzipieren und umzusetzen.
In Deutschland leidet ca. jeder zehnte Mensch über 40 Jahren an einer chronischen Einschränkung seiner Nierenfunktion. Nicht wenige davon sind im Laufe der Erkrankung auf eine Nierenersatztherapie angewiesen. Die Ursachen für eine Nierenschädigung sind vielfältig. Als neuartiger und vielversprechender Therapieansatz werden aktuell Mesenchymale Stamm-/Stromazellen (MSC) als Therapeutikum für diverse Nierenerkrankungen getestet. Erste Ergebnisse klinischer Phase-I-Studien zeigen, dass MSC sicher als Immunsuppressivum nach Nierentransplantation angewendet werden können. Auch für weitere Erkrankungen der Niere sind erste klinische Studien am Laufen. MSC gelten als regenerativ, immunsupprimierend und antientzündlich. Dennoch gibt es noch einige Limitationen. Nach der Transplantation der Zellen ist das Wachstum der Zellen oft eingeschränkt und es kommt zur vermehrten Apoptose. Auch wird immer wieder ein paradoxes und entzündungsförderndes Verhalten der MSC am Wirkort beobachtet. Ein wichtiger Lösungsansatz ist eine in vitro Vorbehandlung der MSC zur Modulierung der zellulären Eigenschaften. In dieser Arbeit wurden drei Substanzen und Arzneimittel auf ihre Fähigkeit untersucht, die entzündungshemmenden Eigenschaften der MSC zu verbessern und die entzündungsfördernden zu unterdrücken. Der Fokus lag hierbei auf dem Arzneimittel Niclosamid und den beiden bisher noch nicht zugelassenen Substanzen Berberin und Gedunin, die alle in vitro und in vivo bereits erste vielversprechende antientzündliche Wirkungen bewiesen haben. Für diese Arbeit wurden MSC aus Fettgewebe isoliert (ASC) und mit LPS oder einem Cytokin-Mix (Mischung aus TNF-α, IFN-γ und IL-1β) sowie den drei Substanzen stimuliert. Untersucht wurden im Anschluss die mRNA-Expressionen der gängigsten proinflammatorischen (TNF-α, IL-6, TLR-4, ICAM-1, HLA-DR) und antiinflammatorischen Marker (IDO, IL-10), sowie mittels ELISA die Protein-Freisetzung von IL-6 und IL-8. Die vielversprechendsten Ergebnisse ließen sich durch Berberin hervorrufen. Hier zeigte sich eine deutliche Senkung der IL-8-Konzentration im ELISA. Die Anwendung des Gedunin hatte keine signifikante Wirkung auf die ASC. Niclosamid hingegen scheint widererwarten sogar entzündungsfördernd zu wirken, in dem es die IL-6-, ICAM-1-mRNA-Expression steigerte und die IDO-mRNA-Expression absenkte. Unter den drei getesteten Subtanzen hat Berberin die beste Wirkung bewiesen. Nach weiterer Testung könnte eine Anwendung mit Berberin als in vitro Präkonditionierung von MSC vielversprechend sein. Die Verwendung von Niclosamid hingegen sollte vermieden werden, die Wirkung von Gedunin müsste genauer untersucht werden.
Hidradenitis suppurativa ist eine multifaktoriell-bedingte chronisch entzündliche Hauterkrankung, die durch eine Okklusion der Talgdrüseneinheit des Haarfollikels entsteht. Aus der anschließend mit Entzündung einhergehenden Ruptur des Haarfollikels entwickeln sich entzündliche Knoten, Abszesse und Fistelgänge. (39–41) In der weiteren Progression der Erkrankung kommt es zur Störung der Hautarchitektur und fibrotischen Narbenbildungen. (52) Durch Untersuchungen des entzündlichen Infiltrates konnte bereits die Beteiligung einer Reihe von Immunzellen und Entzündungsmediatoren identifiziert werden. Hierzu zählen Makrophagen, neutrophile Granulozyten, Dendritische Zellen, Lymphozyten, IL-1β sowie TNF-α. (41,45,46,51,53) Da die genaue Pathophysiologie der Hidradenitis suppurativa bislang unzureichend aufgeklärt ist, gibt es aktuell keine kausale Therapiemöglichkeit für die Betroffenen. (52) Die Wahl der Therapie wird anhand der Bewertung des Schweregrades nach Hurley getroffen. (25) Obwohl meisten Patientinnen und Patienten von der milden bis mittelschweren Form der Hidradenitis suppurativa (Hurley I und Hurley II) betroffen sind (98), werden die meisten Arzneimittel für die Behandlung von Hurley II und Hurley III von den Leitlinien empfohlen. Zu den empfohlenen Medikamenten gehören u. a. Rifampicin, meist in Kombination mit Clindamycin, sowie der TNF-α-Inhibitor Adalimumab (18,85), welche effektiv und systemisch wirken. Durch die entstehenden Nebenwirkungen wäre die Behandlung der milden bis mittelschweren Hidradenitis suppurativa mit diesen Medikamenten allerdings unverhältnismäßig. Um jedoch den im Verlauf der Krankheit entstehenden Hautdestruktionen vorbeugen zu können, müssten die Medikamente möglichst früh eingesetzt werden. (228) Vor diesem Hintergrund sollte in Kooperation mit dem Institut für Pharmazeutische Technologie der Goethe Universität Frankfurt eine Rifampicin-Nanoformulierung zur Behandlung milder bis mittelschwerer Hidradenitis suppurativa entwickelt und im Labor der dermatologischen Klinik präklinisch validiert werden.
Daher wurde im ersten Teil der vorliegenden Arbeit ein Epidermismodell aus der Haut betroffener Patientinnen und Patienten generiert, um die Rifampicin-Nanoformulierung validieren zu können. Dieses wies eine mehrschichtige Epidermis mit allen wichtigen Differenzierungsmarkern ähnlich der läsionalen Hidradenitis suppurativa auf und schüttete die proinflammatorischen Zytokine IL-1β und TNF-α aus. Als weiteres Modell wurden ex vivo Explantate aus läsionaler Hidradenitis suppurativa Haut etabliert. Für die Bewertung der Validität der Explantatkulturen wurde die Morphologie und Integrität der Epidermis mittels Hämatoxylin-Eosin sowie der Proliferationsmarker Ki-67 näher beleuchtet. Im zweiten Teil dieser Arbeit wurden Untersuchungen für die Festlegung eines Konzentrationsbereichs, unter Verwendung von Rifampicin in DMSO gelöst als in vitro Behandlung, durchgeführt.
Hierbei wurde gezeigt, dass die eingesetzten Konzentrationen keine negativen Effekte bezüglich Proliferationsfähigkeit der Keratinozyten oder Apoptoseinduktion ausüben. Die Behandlung von nicht entzündlichen Epidermismodellen sowie Explantatkulturen mit der Rifampicin-Nanoformulierung mit einem Wirkstoffgehalt von 0,3 % führte ebenfalls zu keinen Proliferationsverlusten, induzierte keine Apoptose oder Zytotoxizität und hatte keinen Einfluss auf die Differenzierung der Keratinozyten. Im letzten Teil der Arbeit sollte die Wirksamkeit der Rifampicin-Nanoformulierung näher beleuchtet werden. Als Antibiotikum inhibiert Rifampicin die DNA-abhängige RNA-Polymerase von Bakterien (107,108), weshalb es bei der Behandlung der Tuberkulose eingesetzt wird. (111–113) Die Hidradenitis suppurativa ist aber primär keine Infektionskrankheit, sondern eine von Bakterien getriggerte, entzündliche Erkrankung. (19,52,82) Aus diesem Grund ist der positive Effekt der systemischen Rifampicintherapie vermutlich vielmehr auf eine antiphlogistische Wirkung zurückzuführen. Diese Überlegung wird durch mehrere Artikel gestützt, die zeigten, dass Rifampicin die Ausschüttung von IL-1β und TNF-α in unterschiedlichen in vitro und in vivo Modellen hemmt. (89–91,96,97) Die anschließenden Untersuchungen zur Wirksamkeit einer in vitro Rifampicinbehandlung bestätigten die antientzündliche Wirkung in den Explantatkulturen indem es die Sekretion von IL-1β, IL-6, IL-8, IL-10 und TNF-α verminderte. Ebenso senkte die Rifampicin-Nanoformulierung die Ausschüttung von IL-1β in den ex vivo Explantaten, was somit die in der klinischen Praxis beobachteten antiinflammatorischen Wirkung von Rifampicin belegt. Des Weiteren stellte sich in Untersuchungen heraus, dass Rifampicin zu einer reduzierten Zahl CD4(+)-T-Zellen führte, aber auf die CD3(+)-T-Zellen keine Auswirkungen hatte, was auf eine Veränderung des T-Zell Phänotyps hinweist.
Aufgrund der vorliegenden Ergebnisse zur Validierung der Rifampicin-Nanoformulierung, spricht nichts gegen die Testung der Rifampicin-Nanoformulierung in einem individuellen Heilversuch am Menschen.
We report results on an elastic cross section measurement in proton–proton collisions at a center-of-mass energy √𝑠 = 510 GeV, obtained with the Roman Pot setup of the STAR experiment at the Relativistic Heavy Ion Collider (RHIC). The elastic differential cross section is measured in the four-momentum transfer squared range 0.23 ≤ −𝑡 ≤ 0.67 GeV2. This is the only measurement of the proton-proton elastic cross section in this 𝑡 range for collision energies above the Intersecting Storage Rings (ISR) and below the Large Hadron Collider (LHC) colliders. We find that a constant slope 𝐵 does not fit the data in the aforementioned 𝑡 range, and we obtain a much better fit using a second-order polynomial for 𝐵(𝑡). This is the first measurement below the LHC energies for which the non-constant behavior 𝐵(𝑡) is observed. The 𝑡 dependence of 𝐵 is also determined using six subintervals of 𝑡 in the STAR measured 𝑡 range, and is in good agreement with the phenomenological models. The measured elastic differential cross section d𝜎∕dt agrees well with the results obtained at √𝑠 = 540 GeV for proton–antiproton collisions by the UA4 experiment. We also determine that the integrated elastic cross section within the STAR 𝑡-range is 𝜎f id el = 462.1 ± 0.9(stat.) ± 1.1(syst.) ± 11.6(scale) 𝜇b.
Particle production as a function of charged-particle flattenicity in pp collisions at √s = 13 TeV

This paper reports the first measurement of the transverse momentum (pT) spectra of primary charged pions, kaons, (anti)protons, and unidentified particles as a function of the charged-particle flattenicity in pp collisions at s√=13 TeV. Flattenicity is a novel event shape observable that is measured in the pseudorapidity intervals covered by the V0 detector, 2.8<η<5.1 and −3.7<η<−1.7. According to QCD-inspired phenomenological models, it shows sensitivity to multiparton interactions and is less affected by biases towards larger pT due to local multiplicity fluctuations in the V0 acceptance than multiplicity. The analysis is performed in minimum-bias (MB) as well as in high-multiplicity events up to pT=20 GeV/c. The event selection requires at least one charged particle produced in the pseudorapidity interval |η|<1. The measured pT distributions, average pT, kaon-to-pion and proton-to-pion particle ratios, presented in this paper, are compared to model calculations using PYTHIA 8 based on color strings and EPOS LHC. The modification of the pT-spectral shapes in low-flattenicity events that have large event activity with respect to those measured in MB events develops a pronounced peak at intermediate pT (2<pT<8 GeV/c), and approaches the vicinity of unity at higher pT. The results are qualitatively described by PYTHIA, and they show different behavior than those measured as a function of charged-particle multiplicity based on the V0M estimator.
Measurement of beauty production via non-prompt charm hadrons in p-Pb collisions at √sNN = 5.02 TeV

The production cross sections of D0, D+, and Λ+c hadrons originating from beauty-hadron decays (i.e. non-prompt) were measured for the first time at midrapidity in proton−lead (p−Pb) collisions at the center-of-mass energy per nucleon pair of √sNN=5.02 TeV. Nuclear modification factors (RpPb) of non-prompt D0, D+, and Λ+c are calculated as a function of the transverse momentum (pT) to investigate the modification of the momentum spectra measured in p−Pb collisions with respect to those measured in proton−proton (pp) collisions at the same energy. The RpPb measurements are compatible with unity and with the measurements in the prompt charm sector, and do not show a significant pT dependence. The pT-integrated cross sections and pT-integrated RpPb of non-prompt D0 and D+ mesons are also computed by extrapolating the visible cross sections down to pT = 0. The non-prompt D-meson RpPb integrated over pT is compatible with unity and with model calculations implementing modification of the parton distribution functions of nucleons bound in nuclei with respect to free nucleons. The non-prompt Λ+c/D0 and D+/D0 production ratios are computed to investigate hadronisation mechanisms of beauty quarks into mesons and baryons. The measured ratios as a function of pT display a similar trend to that measured for charm hadrons in the same collision system.
The production yields of antideuterons and antiprotons are measured in pp collisions at a center-of-mass energy of √s=13 TeV, as a function of transverse momentum (pT) and rapidity (y), for the first time up to |y|=0.7. The measured spectra are used to study the pT and rapidity dependence of the coalescence parameter B2, which quantifies the coalescence probability of antideuterons. The pT and rapidity dependence of the obtained B2 is extrapolated for pT>1.7 GeV/c and |y|>0.7 using the phenomenological antideuteron production model implemented in PYTHIA 8.3 as well as a baryon coalescence afterburner model based on EPOS 3. Such measurements are of interest to the astrophysics community, since they can be used for the calculation of the flux of antinuclei from cosmic rays, in combination with coalescence models.
Neurodevelopmental psychiatric disorders (NPDs) like attention deficit hyperactivity disorder (ADHD), autism spectrum disorder (ASD), and schizophrenia, affect millions of people worldwide. Despite recent progress in NPD research, much remains to be discovered about their underpinnings, therapeutic targets, effects of biological sex and age. Risk factors influencing brain development and signalling include prenatal inflammation and genetic variation. This dissertation aimed to build upon these findings by combining behavioural, molecular, and neuromorphological investigations in mouse models of such risk factors, i.e. maternal immune activation (MIA), neuron-specific overexpression (OE) of the cytoplasmatic isoforms of the RNA-binding protein RBFOX1, and neuronal deletion of the small Ras GTPase DIRAS2.
Maternal infections during pregnancy pose an increased risk for NPDs in the offspring. While viral-like MIA has been previously established elsewhere, this study was the first in our institution to implement the model. I validated NPD-relevant deficits in anxiety- and depression-like behaviours, as well as dose- and sex-specific social deficits in mouse offspring following MIA in early gestation. Proteomic analyses in embryonic and adult hippocampal (HPC) synaptoneurosomes highlighted novel and known targets affected by MIA. Analysis of the embryonic dataset implicated neurodevelopmental disruptions of the lipid, polysaccharide, and glycoprotein metabolism, important for proper membrane function, signalling, and myelination, for NPD-pertinent sequelae. In adulthood, the observed changes encompassed transmembrane trafficking and intracellular signalling, apoptosis, and cytoskeletal organisation pathways. Importantly, 50 proteins altered by MIA in embryonic and adult HPC were enriched in the NPD-relevant synaptic vesicle cycle. A persistently upregulated protein cluster formed a functional network involved in presynaptic signalling and proteins downregulated in embryos but upregulated in adults by MIA were correlated with observed social deficits. 49/50 genes encoding these proteins were significantly associated with NPD- and comorbidity-relevant traits in human phenome-wise association study data for psychiatric phenotypes. These findings highlight NPD-relevant targets for future study and early intervention in at-risk individuals. MIA-evoked changes in the neuroarchitecture of the NPD-relevant HPC and prefrontal cortex (PFC) of male and female mice highlighted sex- and region-specific alterations in dendritic and spine morphology, possibly underlining behavioural phenotypes.
To further investigate genetic risk factors of NPDs, I performed a study based on the implications of RBFOX1’s pleiotropic role in neuropsychiatric disorders and previous preclinical findings. Cytoplasmatic OE of RBFOX1, which affects the stability and translation of thousands of targets, was used to disseminate its role in morphology and behaviour. RBFOX1 OE affected dendritic length and branching in the male PFC and led to spine alterations in both PFC and HPC. Due to previously observed ASD-like endophenotypes in our Rbfox1 KO mice and the importance of gene × environment effects on NPD susceptibility, I probed the interaction of cytoplasmatic OE and a low-dose MIA on offspring. Both RBFOX1 OE alone and with MIA led to increased offspring loss during the perinatal period. Preliminary data suggested that RBFOX1 OE × MIA might increase anxiety- and anhedonia-like behaviours. Morphological changes in the adult male OE HPC and PFC suggested increased spine density and reduced dendritic complexity. A small post-mortem study in human dorsolateral PFC of older adults did not reveal significant effects of a common risk variant on RBFOX1 abundance.
To expand upon NPD genetic risks, I evaluated the effects of a homo- (KO) or heterozygous (HET) Diras2 deletion in a novel, neuron-specific mouse model. DIRAS2’s function is largely unknown, but it has been associated with ADHD in humans and neurodevelopment in vitro. In adult mice, there were subtle sex-specific effects on behaviour, i.e. more pronounced NPD-relevant deficits in males, in keeping with human data. KO mice had subtly improved cognitive performance, while HET mice exhibited behaviours in line with core ADHD symptoms, e.g. earning difficulties (females), response inhibition deficits and hyperactivity (males), suggesting Diras2 dose-sensitivity and sex-specificity. The morphological findings revealed multiple aberrations in dendritic and spine morphology in the adult PFC, HPC, and amygdala of HET males. KOs changes in spine and dendritic morphology were exclusively in the PFC and largely opposite to those in HETs and NPD-like phenotypes. Region- and genotype-specific expression changes in Diras2 and Diras1 were observed in six relevant brain regions of adult HET and KO females, also revealing differences in the survival and morphology regulator mTOR, which might underlie observed differences.
In conclusion, the effects of MIA and partial Diras2 knockdown resembled each other in core, NPD-associated behavioural and morphological phenotypes, while cytoplasmatic RBFOX1 OE and full Diras2 KO differed from those. My findings suggest complex dose- and sex-dependent relationships between these prenatal and genetic interventions, whose NPD-relevant influences might converge onto neurodevelopmental molecular pathways. An assessment of such putative overlap, based on available data from the MIA proteomic analyses of embryonic and adult HPC, suggested the three models might be linked via downstream targets, interactions, and upstream regulators. Future studies should disseminate both distinct and shared aspects of MIA, RBFOX1, and DIRAS2 relevant to NPDs and build upon these findings.
Exploring strategies to improve the reverse beta-oxidation pathway in Saccharomyces cerevisiae

Microbes are the most diverse living organisms on Earth, with various metabolic adaptations that allow them to live in different conditions and produce compounds with different chemical complexity. Microbial biotechnology exploits the metabolic diversity of microorganisms to manufacture products for different industries. Today, the chemical industry is a significant energy consumer and carbon dioxide emitter, with processes that harm natural ecosystems, like the extraction of medium-chain fatty acids (MCFAs). MCFAs are used as precursors for biofuels, volatile esters, surfactants, or polymers in materials with enhanced properties.
However, their current extraction process uses large, non-sustainable monocultures of coconut and palm trees. Therefore, the microbial production of MCFAs can help reduce the current environmental impact of obtaining these products and their derivatives.
In nature, fatty acids are mostly produced via fatty acid biosynthesis (FAB). However, the reverse β-oxidation (rBOX) is a more energy-efficient pathway compared to FAB. The rBOX pathway consists of four reactions, which result in the elongation of an acyl-CoA molecule by two carbon units from acetyl-CoA in each cycle. In this work we used Saccharomyces cerevisiae, an organism with a high tolerance towards toxic compounds, as the expression host of the rBOX pathway to produce MCFAs and medium-chain fatty alcohols (MCFOHs).
In the first part of this work, we expanded the length of the products from expressing the rBOX in the cytosol and increased the MCFAs titres. First, we deleted the major glycerol-3-phosphate dehydrogenase (GPD2). This resulted in a platform strain with significantly reduced glycerol fermentation and increased rBOX pathway activity, probably due to an increased availability of NADH. Then, we tested different combinations of rBOX enzymes to increase the length and titres of MCFA. Expressing the thiolase CnbktB and β-hydroxyacyl-CoA dehydrogenase CnpaaH1 from Cupriavidus necator, Cacrt from Clostridium acetobutylicum and the trans-enoyl-CoA reductase Tdter (Treponema denticola) resulted in hexanoic acid as the main product.
Expressing Cncrt2 (C. necator) or YlECH (Y. lipolytica) as enoyl-CoA hydratases resulted in octanoic acid as the main product. Then, we integrated the octanoic (Cncrt2 or YlECH) and the hexanoic acid (Cacrt)-producing variants in the genome of the platform strain and we achieved titers of ≈75 mg/L (hexanoic acid) and ≈ 60 mg/L (octanoic acid) when growing these strains in a complex, highly buffered medium. These are the highest titers of octanoic and hexanoic acid obtained in S. cerevisiae with the rBOX. Additionally, we deleted TES1 and FAA2 to prevent competition for butyryl-CoA and degradation of the produced fatty acids, respectively.
However, these deletions did not improve MCFA titers. In addition, we tested two dual acyl-CoA reductase/alcohol dehydrogenases (ACR/ADH), CaadhE2 from C. acetobutylicum and the putative ACR/ADH EceutE from Escherichia coli, in an octanoyl-CoA-producing strain to produce MCFOH. As a result, we produced 1-hexanol and 1-octanol for the first time in S. cerevisiae with these two enzymes. Nonetheless, the titres were low (<10 mg/L and <2 mg/L, respectively), and four-carbon 1-butanol was the main product in both cases (>80 mg/L). This showed the preference of these two enzymes for butyryl-CoA.
In the second part of this work, we expressed the rBOX in the mitochondria of S. cerevisiae to benefit from the high levels of acetyl-CoA and the reducing environment in that organelle. First, in an adh-deficient strain, we mutated MTH1, a transcription factor regulating the expression of hexose transporters, and deleted GPD2. This resulted in a strain with a reduced Crabtree effect and, therefore, an increased carbon flux to the mitochondria. We partially validated the increased flux to the mitochondria by expressing the ethanol-acetyltransferase EAT1 from Kluyveromyces marxianus in this organelle. This resulted in a higher isoamyl acetate production in the MTH1-mutant strain. Isoamyl acetate is synthesised by Eat1 from acetyl-CoA and isoamyl alcohol, a product of the metabolism of amino acids in the mitochondria. Then, we targeted different butyryl-CoA-producing rBOX variants to the mitochondria, and we used the production of 1-butanol and butyric acid as a proof-of-concept. The strong expression of all the enzymes was toxic for the cell, and the highest butyric acid titres (≈ 50 mg/L) in the mitochondria from the rBOX were obtained from the weak expression of the pathway. The highest 1-butanol titers (≈ 5 mg/L) were obtained with the downregulation of the mitochondrial NADH-oxidase NDI1. However, this downregulation led to a non-desirable petite phenotype.
In summary, we produced hexanoic and octanoic acid for the first time in S. cerevisiae using the rBOX and achieved the highest reported titers of hexanoic and octanoic acid so far using this pathway in S. cerevisiae. In addition, we successfully compartmentalised the rBOX in the mitochondria. However, competing reactions, some of them essential for the viability of the cell, limit the use of this organelle for the rBOX.
Research on the human and animal microbiome has become increasingly important in recent years. It is now widely accepted the gut microbiome is of crucial importance to health, as it is involved in a large number of physiological processes. The term ‘microbiome’ refers to the all living microorganisms including their genes and metabolites in a defined environment, while the specific composition of microorganisms consisting of bacteria, archaea and protozoa is referred to as the ‘microbiota’ (Lane-Petter, 1962; Lederberg and McCray, 2001).
In recent years, research has focused on various of these communities in the soil (Fierer, 2017), water (Sunagawa et al., 2015), air (Leung et al., 2014) and especially in the human gut. However, this topic is also becoming increasingly relevant for the conservation of endangered species. In the face of global mass extinctions and the listing of over 42,000 animal species as ‘critically endangered’, conservation breeding programmes are more important than ever (Díaz et al., 2019; IUCN, 2022). The responsibility for these tasks lies with zoological institutions, which are dedicated to animal conservation and the continuous monitoring of animal welfare. Microbiome research offers a non-invasive method to support species conservation. By analysing faecal samples, microbial markers can be identified that provide important information about the health status and reproductive cycle of animals (Weingrill et al., 2004; Antwis et al., 2019). Zoological facilities also provide an ideal research environment for comparing individuals from different habitats. In addition, all necessary metadata such as age, sex, kinship or medical treatment are documented and can be used for the analysis.
This is the starting point for this thesis. In order to identify such microbial markers, it is necessary to understand the microbiome of a variety of animal species. The first aim is therefore to characterise the faecal microbiota of 31 mammalian species, focusing on herbivores and carnivores. It could be shown that they differ significantly in terms of both microbial diversity and microbiota composition. Herbivorous species express a very diverse microbial composition, consisting mainly of cellulose-degrading taxa of the families Fibrobacteraceae or Spirochaetaceae. In contrast, the microbiota of carnivorous species is less diverse and is dominated by protein-degrading Fusobacteriaceae and Clostridiaceae. In addition, this thesis proves that the microbiota of herbivorous species is highly consistent, whereas the microbiota of carnivorous species is highly variable. The results of this study provide important insights for the sampling scheme of future projects. Especially when analysing carnivorous species, single samples are not sufficient to capture the full variability of the microbiome.
These results lead to the question of whether this variability can be explained by daily fluctuations in the individual microbiome and whether this can be used to distinguish between species or individuals. Using individual longitudinal data and a combined approach of clustering algorithms and dynamic time warping, it is shown that such a distinction is possible at the species and individual level. This was confirmed for both a carnivorous (Panthera tigris) and a herbivorous (Connochaetes taurinus) species. These results confirm the influence of the host individual on the faecal microbiota, in addition to the often described influence of diet (Ley et al., 2008a; Kartzinel et al., 2019).
Based on the knowledge gained from these studies, a methodology has been developed that will enable the conservation of species in the field to be supported by microbiome research in the future. The focus here lays on the identification of host-specific metadata based on the faecal microbiota. The developed regression model is able to distinguish between carnivorous, herbivorous and omnivorous hosts with up to 99% accuracy. In addition, a more accurate phylogenetic classification of the family (Canidae, Felidae, Ursidae, Herpestidae) can be made for carnivorous hosts. For herbivorous hosts, the model can predict the respective digestive system with up to 100% accuracy, distinguishing between ruminants, hindgut fermenters and a simple digestive system. The acquisition of host-specific metadata from an unknown faecal sample is an important step towards establishing microbiome research in species conservation. Field studies in particular will benefit from such new methods. Usually, costly microsatellite analysis and high-quality host DNA are required to obtain host-specific information from faecal samples. The newly developed method offers a less costly and labour-intensive alternative to conventional techniques and opens up a more accessible field for microbiome research in the field.

Und sowohl Vandana Singhs Erzählung "Of Love and Other Monsters" (2007) als auch Nnedi Okorafors Roman "Lagoon" (2014) erschaffen eine form- und geschlechtswandelnde Spezies von Außerirdischen. Den Erzählungen gelingt es, neue Welten zu entwerfen, die fluide Identitäten reflektieren. Sie geben Anlass dazu, die binären Denkstrukturen unserer Gesellschaft zu hinterfragen und alternative Gesellschaftsformen zu denken. Sie geben außerdem einen Hinweis darauf, in welcher Weise sich der Umgang mit binären Denkstrukturen seit der Veröffentlichung von "The Left Hand of Darkness" entwickelt hat und inwiefern es innerhalb der Literatur und unserer Gesellschaft möglich ist, diese zu transformieren.
Der Hirntumor Glioblastom (GBM) ist aufgrund seines infiltrativen Wachstums, der hohen intra- und intertumoralen Heterogenität, der hohen Therapieresistenz als auch aufgrund der sogenannten gliomartigen Stammzellen sehr schwer zu behandeln und führt fast immer zu Rezidiven. Da es in den letzten Jahrzehnten kaum Fortschritte in der Behandlung des GBMs gab, bis auf die Therapie mit Tumortherapiefeldern, wird weiterhin nach alternativen Zelltodtherapien geforscht, wie zum Beispiel dem Autophagie-abhängigen Zelltod. Der Autophagie-abhängige Zelltod ist durch einen erhöhten autophagischen Flux gekennzeichnet und obwohl die Autophagie, als auch selektive Formen wie die Lysophagie und Mitophagie, normalerweise als überlebensfördernde Mechanismen gelten, konnten viele Studien eine duale Rolle in der Tumorentstehung, -progression und -behandlung aufzeigen, die vor allem vom Tumortyp und stadium abhängt. Um die zugrunde liegenden Mechanismen des durch Medikamente induzierten Autophagie-abhängigen Zelltods im GBM weiter zu entschlüsseln, habe ich in meiner Dissertation verschiedene Substanzen untersucht, die einen Autophagie-abhängigen Zelltod induzieren.
In einer zuvor in unserem Labor durchgeführten Studie konnte gezeigt werden, dass das Antipsychotikum Pimozid (PIMO) und der Opioidrezeptor-Antagonist Loperamid (LOP) einen Autophagie-abhängigen Zelltod in GBM Zellen induzieren können. Darauf aufbauend habe ich die Fähigkeit zur Induktion des Autophagie-abhängigen Zelltods in weiteren Zellmodellen validiert. Dies bestätigte einen erhöhten autophagischen Flux nach PIMO und LOP Behandlung, während der Zelltod als auch der autophagische Flux in Autophagie-defizienten Zellen reduziert war. In weiteren Versuchen konnte ich die Involvierung der LC3-assoziierten Phagozytose (LAP), ein Signalweg der auf die Funktion einiger autophagischer Proteine angewiesen ist, ausschließen. Weiterhin konnte ich eine massive Störung des Cholesterin- und Lipidstoffwechsels beobachten. Unter anderem akkumulierte Cholesterin in den Lysosomen gefolgt von massiven Schäden des lysosomalen Kompartiments und der Permeabiliserung der lysosomalen Membran. Dies trug einerseits zur Aktivierung überlebensfördernder Lysophagie als auch der Zell-schädigenden „Bulk“-Autophagie bei. Letztendlich konnte aber die erhöhte Lysophagie die Zellen nicht vor dem Zelltod retten und die Zellen starben einen Autophagie-abhängigen lysosomalen Zelltod. Da die Eignung von LOP als Therapie für das GBM aufgrund der fehlenden Blut-Hirn-Schranken Permeabilität und von dem Antipsychotikum PIMO aufgrund teils schwerer Nebenwirkungen eingeschränkt ist, habe ich mich im weiteren Verlauf meiner Dissertation mit einer Substanz mit einem anderen Wirkmechanismus beschäftigt.
Der Eisenchelator und oxidative Phosphorylierungs (OXPHOS) Inhibitor VLX600 wurde zuvor berichtet mitochondriale Dysfunktion und Zelltod in Kolonkarzinomzellen zu induzieren. Allerdings hat meines Wissens nach bisher noch keine Studie die therapeutische Eignung von VLX600 für das GBM untersucht. Hier zeige ich eine neuartige Autophagie-abhängige Zelltod-induzierende Fähigkeit von VLX600 für GBM Zellen, da der Zelltod signifikant in Autophagie-defizienten Zellen aber nicht durch Caspase-Inhibitoren gehemmt wurde und der autophagische Flux erhöht war. Darüber hinaus konnte ich die Hemmung der OXPHOS und die Induktion von mitochondrialem Stress in GBM Zellen bestätigen und weiterhin aufzeigen, dass VLX600 nicht nur die mitochondriale Homöostase stört, sondern auch zu einer BNIP3-BNIP3L-abhängigen Mitophagie führt, die wahrscheinlich durch HIF1A reguliert wird aber keinen erkennbaren Nettoeffekt auf den von VLX600 induzierten Zelltod hat. Demnach induziert VLX600 letale „Bulk“-Autophagie in den hier verwendeten Zellmodellen. Darüber hinaus konnte ich zeigen, dass die Eisenchelatierung durch VLX600 eine große Rolle für den von VLX600-induzierten Zelltod spielt aber auch für die Mitophagie Induktion, Histon Lysin Methylierung und den ribosomalen Stress. Letztendlich ist es wahrscheinlich ein Zusammenspiel all dieser Faktoren, die zur Zelltodinduktion durch VLX600 führen und interessanterweise werden Eisenchelatoren bereits in präklinischen und klinischen Studien für Krebstherapien untersucht. Dabei könnten gewisse metabolische Eigenschaften verschiedener Tumorzellen die Sensitivität von Wirkstoffen, die auf den Metabolismus wirken wie VLX600, beeinflussen was in zukünftigen Studien beachtet werden sollte um den bestmöglichsten Therapieerfolg zu erzielen. Zusammenfassend unterstützt meine Dissertation die duale Rolle der Autophagie, die stark vom jeweiligen Kontext abhängt und befürwortet die weitere Forschung von Substanzen, die einen Autophagie-abhängigen Zelltod induzieren, für das GBM.
Der aus Ungarn stammende israelische Satiriker Ephraim Kishon (1924–2005) gilt als ‚Versöhnungsfigur‘ zwischen Deutschen und Jüdinnen und Juden im bundesdeutschen Nachkriegsdiskurs. Seine „israelischen Satiren“ erfreuten sich in der freien Übertragung durch Friedrich Torberg vor allem in den 1960er bis 1990er Jahren enormer Beliebtheit. Dabei wurde zunächst verdrängt, dass Kishon selbst Überlebender der Schoah war und seinen Humor als Überlebensstrategie entwickelt hatte. Bisher wurde die Bedeutung der Schoah für Kishons Schreiben nur unzureichend berücksichtigt.
Birgit M. Körner beleuchtet das Phänomen von Kishons Erfolg in der Bundesrepublik nun von drei Seiten: von der Seite des Autors und Schoah-Überlebenden Kishon, von der Seite des Mitschöpfers und Übersetzers Friedrich Torberg und von der Seite der Rezeption durch ein postnationalsozialistisches deutschsprachiges Publikum.
Im Fokus steht zunächst die Rekonstruktion von Kishons Verfolgungs- und Überlebenserfahrung anhand bisher unbekannter Akten und der Nachweis, dass sich deren Spuren in Kishons Satiren finden lassen. Kishon und Torberg konstruieren einen „israelischen Humor“, der maßgeblich auf den europäischen jüdischen Humortraditionen – dem ostjüdischen Witz und der jüdischen Tradition des literarischen Sarkasmus – sowie auf Kishons Schoah-Überleben basiert. Deutlich wird dabei Torbergs Tendenz, das deutschsprachige Publikum zu ‚schonen‘ und explizite Stellen zu streichen, u.a. um eine positive Haltung zu Israel zu fördern. Kishon selbst stand seiner Rolle als ‚Versöhnungsfigur‘ für ein westdeutsches Publikum durchaus ambivalent gegenüber.
The strong force is one of the four fundamental interactions, and the theory of it is called Quantum Chromodynamics (QCD). A many-body system of strongly interacting particles (QCD matter) can exist in different phases depending on temperature (T) and baryonic chemical potential (µB). The phases and transitions between them can be visualized as µB−T phase diagram. Extraction of the properties of the QCD matter, such as compressibility, viscosity and various susceptibilities, and its Equation of State (EoS) is an important aspect of the QCD matter study. In the region of near-zero baryonic chemical potential and low temperatures the QCD matter degrees of freedom are hadrons, in which quarks and gluons are confined, while at higher temperatures partonic (quarks and gluons) degrees of freedom dominate. This partonic (deconfined) state is called quark-gluon plasma (QGP) and is intensively studied at CERN and BNL. According to lattice QCD calculations at µB=0 the transition to QGP is smooth (cross-over) and takes place at T≈156 MeV. The region of the QCD phase diagram, where matter is compressed to densities of a few times normal nuclear density (µB of several hundreds MeV), is not accessible for the current lattice QCD calculations, and is a subject of intensive research. Some phenomenological models predict a first order phase transition between hadronic and partonic phases in the region of T≲100 MeV and µB≳500 MeV. Search for signs of a possible phase transition and a critical point or clarifying whether the smooth cross-over is continuing in this region are the main goals of the near future explorations of the QCD phase diagram.
In the laboratory a scan of the QCD phase diagram can be performed via heavy-ion collisions. The region of the QCD phase diagram at T≳150 MeV and µB≈0 is accessible in collisions at LHC energies (√sNN of several TeV), while the region of T≲100 MeV and µB≳500 MeV can be studied with collisions at √sNN of a few GeV. The QCD matter created in the overlap region of colliding nuclei (fireball) is rapidly expanding during the collision evolution. In the fireball there are strong temperature and pressure gradients, extreme electromagnetic fields and an exchange of angular momentum and spin between the system constituents. These effects result in various collective phenomena. Pressure gradients and the scattering of particles, together with the initial spatial anisotropy of the density distribution in the fireball, form an anisotropic flow - a momentum (azimuthal) anisotropy in the emission of produced particles. The correlation of particle spin with the angular momentum of colliding nuclei leads to a global polarization of particles. A strong initial magnetic field in the fireball results in a charge dependence and particle-antiparticle difference of flow and polarization.
Anisotropic flow is quantified by the coefficients vₙ from a Fourier decomposition of the azimuthal angle distribution of emitted particles relative to the reaction plane spanned by beam axis and impact parameter direction. The first harmonic coefficient v₁ quantifies the directed flow - preferential particle emission either along or opposite to the impact parameter direction. The v₁ is driven by pressure gradients in the fireball and thus probes the compressibility of the QCD matter. The change of the sign of v₁ at √sNN of several GeV is attributed to a softening of the EoS during the expansion, and thus can be an evidence of the first order phase transition. The global polarization coefficient PH is an average value of the hyperon’s spin projection on the direction of the angular momentum of the colliding system. It probes the dynamics of the QCD matter, such as vorticity, and can shed light on the mechanism of orbital momentum transfer into the spin of produced particles.
In collisions at √sNN of several GeV, which probe the region of the QCD phase diagram at T≲100 MeV and µB≳500 MeV, hadron production is dominated by u and d quarks. Hadrons with strange quarks are produced near the threshold, what makes their yields and dynamics sensitive to the density of the fireball. Thus measurement of flow and polarization, in particular of (multi-)strange particles, provides experimental constraints on the EoS, that allows to extract transport coefficients of the QCD matter from comparison of data with theoretical model calculations of heavy-ion collisions.

In der Welt der wilden Kerle : eine populäre Serie im Zeichen des russisch-ukrainischen Krieges

Zum Jahreswechsel 2023/2024 gelang einer russischen Fernsehserie, was während Russlands Krieg gegen die Ukraine eigentlich unvorstellbar scheint: Innerhalb weniger Tage entwickelte sich "Ehrenwort eines Kerls. Blut auf dem Asphalt" ("Slowo pazana. Krow na asfalte", 2023) beiderseits der Schützengräben zur populärsten Serie des Jahres. Die Zuschauer- und Klickzahlen erreichten Rekordhöhen und der Titelsong "Pyjala" (dt. 'Glas') der tatarischen Band Aigel schaffte es an die Spitze diverser Hitparaden in beiden Ländern. In der Russischen Föderation war die Serie zwar mit der Altersgrenze "18+" versehen und nur bei den privaten Streamingdiensten Wink und START zu sehen. Doch schon während der Ausstrahlung der acht Folgen der ersten Staffel vom 9. November bis 21. Dezember 2023 verbreitete die Serie sich blitzschnell über Telegram und andere digitale Kanäle. Sätze wie "Kerle entschuldigen sich nicht" oder "Denk dran, du bist jetzt ein Kerl, du bist jetzt auf der Straße, und ringsherum sind Feinde" wurden zu geflügelten Worten. Pädagogen und Politikerinnen schlugen Alarm, als in der Presse Berichte auftauchten, die von durch die Serie inspirierten Schlägereien berichteten, und zwar sowohl in Russland als auch in der Ukraine. Bevor die letzten Folgen überhaupt ausgestrahlt worden waren, gab es auf "Ehrenwort eines Kerls" bereits in beiden Ländern ein breites Medienecho, wobei die Kritiken kontrovers ausfielen und von enthusiastischer Begeisterung bis zu hellem Entsetzen und kategorischer Ablehnung reichten. In der Ukraine kreiste die Diskussion vor allem um die Frage, ob die Fernsehserie allein schon deshalb gefährliche Kriegspropaganda sei, weil sie aus dem Feindesland kommt. In Russland erregte die vermeintliche Romantisierung der Verbrecherwelt Anstoß. Manche Kritiker deuteten die Serie aber auch als subversiven Zerrspiegel der militärischen Aggression. Was war das aber für ein populärkulturelles Werk, das für so viel Aufmerksamkeit und Aufregung sorgte?
Heutige Diskussionen um 'Artistic Research' oder 'Investigative Aesthetics' reaktivieren die alte Frage nach den Wissens- und Erkenntnisdimensionen von künstlerischer Praxis. Auch in parallel entstehenden literarischen Texten kommt es zu verstärkten Auseinandersetzungen mit Praktiken und Begriffen außerliterarischer Wissensfelder wie den Medien, dem Recht oder den Wissenschaften. Ebenso lassen sich erhöhte Ansprüche ausmachen, mit dem eigenen literarischen Schreiben eine Art Wissen zu erzeugen. Der Beitrag stellt dies beispielhaft an deutschsprachigen Texten zum Jugoslawien-Krieg in den 1990er Jahren dar und diskutiert die literarische Hinwendung zu Formen des Wissens und Ermittelns als ein den forschenden oder investigativen Künsten verwandtes Phänomen.
Lange hat sich die Feststellung von Subjekten auf den menschlichen Körper gerichtet, auf dessen Spur, besonderes Merkmal, Fingerabdruck oder DNA-Profil. Derweil verliert sich dieser indexikalische Zusammenhang in digitalen Erkennungsverfahren, einerseits indem sie nicht länger ein Indiz auf einen unverwechselbaren Körper beziehen, sondern dessen Bild nunmehr mit den Pixel-Verteilungen in tausenden anderen Bilddateien abgleichen, andererseits weil sie, noch vor der Identifizierung einer Person, überhaupt erst einen "Körper" als solchen registrieren müssen. Damit aber verschieben sich all jene subversiven Praktiken, die den Menschen der Detektion zu entziehen trachten: Statt bloßer Maskierung gilt es jetzt, den Körper selbst zu tilgen - durch technische Verwischungen, Oberflächenmodulationen oder Bewegungsmuster außerhalb der KI-Trainingsraster. Derlei Strategien des Entzugs werden in der zeitgenössischen Videokunst erprobt, etwa in Hito Steyerls "How Not to Be Seen" oder Liam Youngs "Choreographic Camouflage". Damit stellt sich zudem die grundsätzliche Frage nach der Verfügbarkeit und Neuform(at)ierung des Körpers im Digitalen; hier nämlich verwandelt er sich vom Zeichen- und Faktenreservoir in Virtualität und stetes Werden, die ihr politisches Potential auch weit jenseits aller Überwachungstechnologie entfalten.
Das ludische Dispositiv virtueller Investigationen : auf Spurensuchen in interaktiven Krimi-Formaten

In interaktiven Kriminalerzählungen verknüpfen sich Medialität und Ästhetik, wobei Computer als Werkzeuge der Ermittlungsarbeit neue Perspektiven auf polizeiliche Effizienz und agency eröffnen. Narrative Spiele nutzen Technologie und Medien häufig auf innovative und selbstreflexive Weise, um immersive Spielwelten und ludische Herausforderungen für die Spielenden zu schaffen. Dieser Artikel verfolgt die historische Entwicklung von Kriminalspielen anhand einzelner Beispiele. Denn diese weisen bedingt durch ihr Erscheinungsdatum spezifische Formen technischer und ästhetischer Verfahren auf, werfen unterschiedliche Fragen zur Natur von Wissen, Medien und Wahrheit auf und eröffnen unterschiedliche Perspektiven auf die Wechselwirkungen zwischen virtueller und realer Kriminalität sowie auf mögliche Einflüsse auf reale kriminologische Praktiken.
In ihren Investigationen greifen Forensic Architecture auf Crowddaten zurück, die Umweltereignisse sensorisch erfassen und von Nutzer:innen auf digitalen Plattformen geteilt werden. Die Animationsvideos der Forschungsagentur lassen sich als Resultate einer kollektiven Intelligenz reflektieren, bei der datenbezogenen Praktiken der Repräsentation als Aneignungen vorangegangener Operationen mit mobilen Sensormedien 'in the field' erscheinen. Der Beitrag fragt nach den ästhetischen Darstellungsweisen der Ereignisrekonstruktion mittels sensorischer Crowddaten. Am Beispiel der Investigation "The Beirut Port Explosion" (2020) wird gezeigt, wie unterschiedliche, an einer objektiven Darstellung orientierte Visualisierungskonventionen produktiv ineinandergreifen. Um eine kohärente Beweisführung zu gewährleisten, werden die Sensordaten in den Kontext eines Raumsimulationsmodells eingefügt. Das Modell fungiert als Ersatzobjekt für die Stadt Beirut und ist somit konstitutiv für die Virtualisierung der Investigation. Dabei rückt in der ästhetischen Analyse jedoch das zirkuläre Verhältnis zwischen Raummodell und Crowddaten in den Fokus, das vor dem Hintergrund produktionsspezifischer und infrastruktureller Fragestellungen problematisiert wird.
Als Carlo Ginzburg die These formulierte, dass die Geisteswissenschaften wie die Kriminalliteratur im sog. "Indizienparadigma" gründeten, hatte er mit Sherlock Holmes einen Detektiv vor Augen, der persönlich den Tatort besichtigte. Dort erhob er Spuren, kombinierte sie und kam in oftmals ingeniösen, aber auch höchst spekulativen Schlussfolgerungen zur Lösung seines Falls. Vor dem Hintergrund aktueller Entwicklungen in Forschung und Fahndung muss dieses materiell und empirisch grundierte "Indizienparadigma" jedoch einer Revision unterzogen werden. Denn seit der Privatdetektiv von "Kommissar Computer" Konkurrenz bekommen hat, haben sich die Investigationspraktiken grundlegend gewandelt: So können computergestützte Fahndungs- und Aufklärungsmethoden eine Besichtigung des Tatorts ersetzen, während algorithmische Wahrscheinlichkeitsrechnung vergangene wie zukünftige Fälle erhellt. Der vorliegende Sammelband mit Beiträgen aus der Literatur-, Medien- und Designwissenschaft untersucht, inwiefern solche "virtuellen Investigationen" in Literatur und Kunst der Gegenwart eine Revision des Indizienparadigmas einschließen - und inwiefern Begriffe der Virtualität bereits die Investigativarbeit im 19. Jahrhundert prägten
Indizien zu lesen, gehört zu den Plausibilisierungspraktiken in alltäglichen wie wissenschaftlichen Kontexten. Über indexikalische Techniken des Schließens ist Wissen semiologisch wie hermeneutisch dynamisiert. Zentral gilt dies für Disziplinen wie Recht, Medizin, Psychologie, Archäologie, Philologie u.a.m. - sie alle folgen Indizien als eine Art 'Allegorie des Verweisens'. Das Indiz als der Zeichentypus, der sich selbst zeigt und zugleich etwas anderes an-zeigt (indicare), changiert dabei stets zwischen Evidenz und Lektüre - das weiß bereits die Antike, doch epistemologisch und semiologisch prominent diskutiert wird es im 18. Jahrhundert.
Improved measurement of the branching fraction of h_(c) → γη^(′)/η and search for h_(c) → γπ⁰

The processes hc→γP(P=η′, η, π0)) are studied with a sample of (27.12±0.14)×108 ψ(3686) events collected by the BESIII detector at the BEPCII collider. The branching fractions of hc→γη′ and hc→γη are measured to be (1.40±0.11±0.04±0.10)×10−3 and (3.77±0.55±0.13±0.26)×10−4, respectively, where the first uncertainties are statistical, the second systematic, and the third from the branching fraction of ψ(3686)→π0hc. The ratio Rhc=B(hc→γη)B(hc→γη′) is calculated to be (27.0±4.4±1.0)%. The measurements are consistent with the previous results with improved precision by a factor of 2. The results are valuable for gaining a deeper understanding of η−η′ mixing, and its manifestation within quantum chromodynamics. No significant signal is found for the decay hc→γπ0, and an upper limit is placed on its branching fraction of B(hc→γπ0)<5.0×10−5, at the 90\% confidence level.
Using data samples collected with the BESIII detector operating at the BEPCII storage ring, the cross section of the inclusive process e+e−→η+X, normalized by the total cross section of e+e−→hadrons, is measured at eight center-of-mass energy points from 2.0000 GeV to 3.6710 GeV. These are the first measurements with momentum dependence in this energy region. Our measurement shows a significant discrepancy from calculations with the existing fragmentation functions. To address this discrepancy, a new QCD analysis is performed at the next-to-next-to-leading order with hadron mass corrections and higher twist effects, which can explain both the established high-energy data and our measurements reasonably well.
The absolute branching fraction of the decay Λc(2625)+→Λ+cπ+π− is measured for the first time to be (50.7±5.0stat.±4.9syst.)% with 368.48 pb−1 of e+e− collision data collected by the BESIII detector at the center-of-mass energies of s√=4.918 and 4.950 GeV. This result is lower than the naive prediction of 67\%, obtained from isospin symmetry, by more than 2σ, thereby indicating that the novel mechanism referred to as the \textit{threshold effect}, proposed for the strong decays of Λc(2595)+, also applies to Λc(2625)+. This measurement is necessary to obtain the coupling constants for the transitions between s-wave and p-wave charmed baryons in heavy hadron chiral perturbation theory. In addition, we search for the decay Λc(2595)+→Λ+cπ+π−. No significant signal is observed, and the upper limit on its branching fraction is determined to be 80.8\% at the 90\% confidence level.
The Cabbibo-favored decay Λ+c→Ξ0K+π0 is studied for the first time using 6.1 fb−1 of e+e− collision data at center-of-mass energies between 4.600 and 4.840 GeV, collected with the BESIII detector at the BEPCII collider. With a double-tag method, the branching fraction of the three-body decay Λ+c→Ξ0K+π0 is measured to be (7.79±1.46±0.71)×10−3, where the first and second uncertainties are statistical and systematic, respectively. The branching fraction of the two-body decay Λ+c→Ξ(1530)0K+ is (5.99±1.04±0.29)×10−3, which is consistent with the previous result of (5.02±0.99±0.31)×10−3. In addition, the upper limit on the branching fraction of the doubly Cabbibo-suppressed decay Λ+c→nK+π0 is 7.1×10−4 at the 90% confidence level. The upper limits on the branching fractions of Λ+c→Σ0K+π0 and ΛK+π0 are also determined to be 1.8×10−3 and 2.0×10−3, respectively.
Evidence for the singly Cabibbo suppressed decay Λ+c→pπ0 is reported for the first time with a statistical significance of 3.7σ based on 6.0 fb−1 of e+e− collision data collected at center-of-mass energies between 4.600 and 4.843 GeV with the BESIII detector at the BEPCII collider. The absolute branching fraction of Λ+c→pπ0 is measured to be (1.56+0.72−0.58±0.20)×10−4. Combining with the branching fraction of Λ+c→nπ+, (6.6±1.3)×10−4, the ratio of the branching fractions of Λ+c→nπ+ and Λ+c→pπ0 is calculated to be 3.2+2.2−1.2. As an important input for the theoretical models describing the decay mechanisms of charmed baryons, our result indicates that the non-factorizable contributions play an essential role and their interference with the factorizable contributions should not be significant. In addition, the absolute branching fraction of Λ+c→pη is measured to be (1.63±0.31stat±0.11syst)×10−3.
We search for the di-photon decay of a light pseudoscalar axion-like particle, a, in radiative decays of the J/ψ, using 10 billion J/ψ events collected with the BESIII detector. We find no evidence of a narrow resonance and set upper limits at the 95% confidence level on the product branching fraction B(J/ψ→γa)×B(a→γγ) and the axion-like particle photon coupling constant gaγγ in the ranges of (3.6−49.8)×10−8 and (2.2−103.8)×10−4 GeV−1, respectively, for 0.18≤ma≤2.85 GeV/c2. These are the most stringent limits to date in this mass region.
Muller's ratchet, in its prototype version, models a haploid, asexual population whose size~N is constant over the generations. Slightly deleterious mutations are acquired along the lineages at a constant rate, and individuals carrying less mutations have a selective advantage. The classical variant considers {\it fitness proportional} selection, but other fitness schemes are conceivable as well. Inspired by the work of Etheridge et al. ([EPW09]) we propose a parameter scaling which fits well to the ``near-critical'' regime that was in the focus of [EPW09] (and in which the mutation-selection ratio diverges logarithmically as N→∞). Using a Moran model, we investigate the``rule of thumb'' given in [EPW09] for the click rate of the ``classical ratchet'' by putting it into the context of new results on the long-time evolution of the size of the best class of the ratchet with (binary) tournament selection, which (other than that of the classical ratchet) follows an autonomous dynamics up to the time of its extinction. In [GSW23] it was discovered that the tournament ratchet has a hierarchy of dual processes which can be constructed on top of an Ancestral Selection graph with a Poisson decoration. For a regime in which the mutation/selection-ratio remains bounded away from 1, this was used in [GSW23] to reveal the asymptotics of the click rates as well as that of the type frequency profile between clicks. We will describe how these ideas can be extended to the near-critical regime in which the mutation-selection ratio of the tournament ratchet converges to 1 as N→∞.
