Fluorescence microscopy has significantly impacted our understanding of cell biology. The extension of diffraction-unlimited super-resolution microscopy opened an observation window that allows for the scrutiny of cellular organization at a molecular level. The non-invasive nature of visible light in super-resolution microscopy methods renders them suitable for observations in living cells and organisms. Building upon these advancements, a promising synergy between super-resolution fluorescence microscopy and deep learning becomes evident, extending the capabilities of the imaging methods. Tasks such as image modality translation, restoration, single-molecule fitting, virtual labeling, spectral demixing, and molecular counting, are enabled with high precision. The techniques explored in this thesis address three critical facets in advanced microscopy, namely the reduction in image acquisition time, saving photon budget during measurement, and increasing the multiplexing capability. Furthermore, descriptors of protein distributions and their motion on cell membranes were developed.
Dass das ZfL ihn seit Kurzem als Senior Fellow führt, passt so gar nicht zu der ungestümen Neugierde und den kreativen Energien, die sich Detlev Schöttker nicht nur bewahrt hat, sondern die jüngst über der Beschäftigung mit einem neuen Gegenstand eine neue Qualität gewonnen haben. Instantan, vehement und bedingungslos hat er sich nach dem Umzug des ZfL nach Wilmersdorf in ein neues Forschungsprojekt weniger vertieft als gestürzt. Doch recht besehen ist es kein neuer Gegenstand, sondern es sind seine alten Bekannten, die ihm rund um den Fasanenplatz wiederbegegnen. Ein größeres Geschenk als diese Nachbarschaft hätte man ihm vielleicht nicht machen können: Die literarische und kulturelle Moderne entstand hier! Der Fasanenplatz ist ein 'Freilichtmuseum' der Moderne mit Hauptmann, Brecht, Benjamin und vielen anderen. Und Detlev Schöttker wäre nicht Detlev Schöttker, wenn er die Öffentlichkeit nicht sogleich über einige seiner Funde und Entdeckungen informiert hätte. In der FAZ sind bereits mehrere Artikel von ihm über die erstaunliche Bedeutung unseres Kiezes für die Moderne erschienen.
Motivated by the on-going discussion on the nature of magnetism in the quantum Ising chain CoNb2O6, we present a first-principles-based analysis of its exchange interactions by applying an \textit{ab initio} approach with additional modelling that accounts for various drawbacks of a purely density functional theory ansatz. With this method we are able to extract and understand the origin of the magnetic couplings under inclusion of all symmetry-allowed terms, and to resolve the conflicting model descriptions in CoNb2O6. We find that the twisted Kitaev chain and the transverse-field ferromagnetic Ising chain views are mutually compatible, although additional off-diagonal exchanges are necessary to provide a complete picture. We show that the dominant exchange interaction is a ligand-centered exchange process - involving the eg electrons -, which is rendered anisotropic by the low-symmetry crystal fields environments in CoNb2O6, giving rise to the dominant Ising exchange, while the smaller bond-dependent anisotropies are found to originate from d−d kinetic exchange processes involving the t2g electrons. We demonstrate the validity of our approach by comparing the predictions of the obtained low-energy model to measured THz and inelastic neutron scattering spectra.
Raumkonstruktionen, die über Handlung und Bedeutungszuschreibungen im Kontext sozialer Medien entstehen, sind ein Fallbeispiel dafür, wie vor dem Hintergrund von Digitalität Fachinhalte re-innoviert werden müssen. Mit dem Ziel eines Beitrags zur Konzeption professionellen Lehrkräftewissens im Kontext von Digitalität werden, ausgehend von der Fragestellung, welche professionellen Fähigkeiten Lehrkräfte benötigen, um Raumkonstruktionen im Unterricht zu thematisieren, in dieser kumulativen Dissertation Raumkonstruktionen aus normativer und empirischer Perspektive als exemplarischer geographischer Fachinhalt adressiert.
Als theoretischer Rahmen dient dabei das TPACK Modell von Mishra & Koehler (2006), das professionelles Lehrkräftewissen in die Bereiche fachliches, pädagogisches und technologisches Wissen sowie deren Überschneidungsbereiche einteilt. Zunächst erfolgt eine Anwendung des Modells als Reflexionsperspektive auf Fachinhalte zur Erzeugung normativer Fähigkeitsbeschreibungen. Diese Fähigkeitsbeschreibungen fließen in die Entwicklung eines TPACK-Selbsteinschätzungsfragebogens für Lehramtsstudierende der Geographie ein. Im Rahmen der damit durchgeführten Studie (n= 364) zeigen sich auf deskriptiver Ebene vergleichsweise niedrige Selbsteinschätzungen der Bereiche inhaltlichen und fachdidaktischen Wissens. Durch die Anwendung einer konfirmatorischen Faktorenanalyse kann das TPACK Modell als zufriedenstellend für die Beschreibung der Daten identifiziert werden. Auffällig ist allerdings die niedrige Korrelation des Konstrukts technologischen Wissens mit den angrenzenden Wissensbereichen. In Bezug auf die Selbsteinschätzungen im phasen- und kontextübergreifenden Vergleich lässt sich, ausgehend von linearen Regressionsanalysen, eine tendenzielle Zunahme der selbsteingeschätzten Fähigkeiten entlang der Fachsemesterzahl ermitteln. Mittels Zweistichproben-t-Tests können außerdem höhere Selbsteinschätzungen der pädagogischen Wissensbereiche durch Studierende, die als Vertretungslehrkräfte tätig sind, festgestellt werden.
In Bezug auf die zur Thematisierung von Raumkonstruktionen im Unterricht benötigten Fähigkeiten ist zunächst die Relevanz der Förderung der Integration fachlichen, pädagogischen und fachdidaktischen Wissens hervorzuheben. Die Studie gibt darüber hinaus Hinweise auf eine niedrigere Bedeutung technologischen Wissens im Hinblick auf soziale Medien als Beispiele alltäglicher Technologien. Vor dem Hintergrund des positiven Effekts von Selbstwirksamkeitsprozessen bieten die Ergebnisse Implikationen für eine Diskussion von praktischen Erfahrungen als Aspekt professionellen Lehrkräftewissens im Sinne der Förderung einer kritisch-reflexiven Auseinandersetzung mit der Tätigkeit als Vertretungslehrkraft aus fachdidaktischer Perspektive. Insgesamt leistet die kumulative Dissertation einen Beitrag zum Diskurs um fachliche geographische Bildung im Kontext von Digitalität und zur Konzeption des professionellen Lehrkräftewissens vor diesem Hintergrund.
Der Dialog ist für die einen das Versprechen gelingender Kommunikation, für die anderen ein überholtes Ideal. Marten Weise zeigt in einer interdisziplinär angelegten Studie, dass sich die Lücke zwischen Lobpreisungen und Abgesängen schließen lässt. Er setzt bei der Unmöglichkeit des Denkens "nach der Shoah" an und erkundet in exemplarischen Untersuchungen der europäischen Literatur-, Theater- und Theoriegeschichte die Spannungen und Widersprüche im Verhältnis zum "Anderen", ohne die der Dialog nicht zu greifen ist. So macht er zwischenmenschliche, soziale und politische Vorgänge als prinzipiell unabschließbares Sprachgeschehen fassbar und eröffnet einen Spielraum für die Aushandlung und das Aushalten von Dissens und Differenz.
The knowledge of the material budget with a high precision is fundamental for measurements of direct photon production using the photon conversion method due to its direct impact on the total systematic uncertainty. Moreover, it influences many aspects of the charged-particle reconstruction performance. In this article, two procedures to determine data-driven corrections to the material-budget description in ALICE simulation software are developed. One is based on the precise knowledge of the gas composition in the Time Projection Chamber. The other is based on the robustness of the ratio between the produced number of photons and charged particles, to a large extent due to the approximate isospin symmetry in the number of produced neutral and charged pions. Both methods are applied to ALICE data allowing for a reduction of the overall material budget systematic uncertainty from 4.5% down to 2.5%. Using these methods, a locally correct material budget is also achieved. The two proposed methods are generic and can be applied to any experiment in a similar fashion.
Memory consolidation tends to be less robust in childhood than adulthood. However, little is known about the corresponding functional differences in the developing brain that may underlie age-related differences in retention of memories over time. This study examined system-level memory consolidation of object-scene associations after learning (immediate delay), one night of sleep (short delay), as well as two weeks (long delay) in 5-to-7-year-old children (n = 49) and in young adults (n = 39), as a reference group with mature consolidation systems. Particularly, we characterized how functional neural activation and reinstatement of neural patterns change over time, assessed by functional magnetic resonance imaging combined with representational similarity analysis (RSA). Our results showed that memory consolidation in children was less robust and strong (i.e., more forgetting) compared to young adults. Contrasting correctly retained remote versus recent memories across time delay, children showed less upregulation in posterior parahippocampal gyrus, lateral occipital cortex, and cerebellum than adults. In addition, both children and adults showed decrease in scene-specific neural reinstatement over time, indicating time-related decay of detailed differentiated memories. At the same time, we observed more generic gist-like neural reinstatement in medial-temporal and prefrontal brain regions uniquely in children, indicating qualitative difference in memory trace in children. Taken together, 5-to-7-year-old children, compared to young adults, show less robust memory consolidation, possibly due to difficulties in engaging in differentiated neural reinstatement in neocortical mnemonic regions during retrieval of remote memories, coupled with relying more on gist-like generic neural reinstatement.
Long- and short-range correlations for pairs of charged particles are studied via two-particle angular correlations in pp collisions at √sNN = 13 TeV and p–Pb collisions at √s = 5.02 TeV. The correlation functions are measured as a function of relative azimuthal angle ∆φ and pseudorapidity separation ∆η for pairs of primary charged particles within the pseudorapidity interval |η| < 0.9 and the transverse-momentum interval 1 < pT < 4 GeV/c. Flow coefficients are extracted for the long-range correlations (1.6 < |∆η| < 1.8) in various high-multiplicity event classes using the low-multiplicity template fit method. The method is used to subtract the enhanced yield of away-side jet fragments in high-multiplicity events. These results show decreasing flow signals toward lower multiplicity events. Furthermore, the flow coefficients for events with hard probes, such as jets or leading particles, do not exhibit any significant changes compared to those obtained from high-multiplicity events without any specific event selection criteria. The results are compared with hydrodynamic-model calculations, and it is found that a better understanding of the initial conditions is necessary to describe the results, particularly for low-multiplicity events.
The production cross section of inclusive isolated photons has been measured by the ALICE experiment at the CERN LHC in pp collisions at centre-of-momentum energy of s√=13 TeV collected during the LHC Run 2 data-taking period. The measurement is performed by combining the measurements of the electromagnetic calorimeter EMCal and the central tracking detectors ITS and TPC, covering a pseudorapidity range of |ηγ|<0.67 and a transverse momentum range of 7<pγT<200 GeV/c. The result extends to lower pγT and xγT=2pγT/s√ ranges, the lowest xγT of any isolated photon measurements to date, extending significantly those measured by the ATLAS and CMS experiments towards lower pγT at the same collision energy with a small overlap between the measurements. The measurement is compared with next-to-leading order perturbative QCD calculations and the results from the ATLAS and CMS experiments as well as with measurements at other collision energies. The measurement and theory prediction are in agreement with each other within the experimental and theoretical uncertainties.
Inorganic phosphate is one of the most abundant and essential nutrients in living organisms. It plays an indispensable role in energy metabolism and serves as a building block for major cellular components such as the backbones of DNA and RNA, headgroups of phospholipids and in posttranslational modifcations of many proteins. Disturbances in cellular phosphate homeostasis have a detrimental effect on the viability of cells. There- fore, both the import and export of phosphate is strictly regulated in eukaryotic cells. In the eukaryotic model organism Saccharomyces cerevisiae, the uptake of phosphate is carried out either by transporters with high affinity or by transporters with low affinity, depending on the cytosolic phosphate concentration. While structures are available for homologues of the high-affinity transporters, no structures of low-affinity transporters have been solved so far. Interestingly, only the low-affinity transporters have a regulatory SPX domain, which is found in various proteins involved in phosphate homeostasis.
In this work, structures of Pho90 from Saccharomyces cerevisiae, a low-affinity phosphate transporter, were solved by cryo-EM, providing insights into its transport mechanism. The dimeric structure resembles the structures of proteins of the divalent anion symporter superfamily (DASS) and of mammalian transporters of the solute carrier 13 (SLC13) family. The transmembrane domain of each protomer consists of 13 helical elements and can be subdivided into scaffold and transport domains. The structure of ScPho90 in the presence of phosphate shows the phosphate binding site within the transporter domain in an outward-open conformation with a bound phosphate ion and two sodium ions. In the absence of phosphate, an asymmetric dimer structure was determined, with one protomer adopting an inward-open conformation. While the dimer contact and the scaffold domain are identical in both conformations, the transport domain is rotated by about 30° and shifted by 11 Å towards the cytoplasmic side, leading to the accessibility of the binding pocket from the cytoplasm. Based on these findings and by comparison with known structures, a phosphate transport mechanism is proposed in the present work that involves substrate binding on the extracellular side, conformational change by a rigid-body motion of the transport domain, in an "elevator-like" motion, and substrate release into the cytoplasm. The regulatory SPX domain is not well resolved in the ScPho90 structures, so that no direct conclusions were drawn about its regulatory mechanism. The findings provide new insights into the function and mechanism of eukaryotic low-affinity phosphate transporters.
While eukaryotic cells express various phosphate import proteins, most eukaryotes have only a single highly conserved and essential phosphate exporter. These exporters show no sequence homology to other transporters of known structure, but also possess a regulatory SPX domain. In this work, the structural basis for eukaryotic phosphate export is investigated by elucidating the structures of the homologous phosphate exporters Syg1 from Saccharomyces cerevisiae and Xpr1 from Homo sapiens, using cryo-EM. The structures of ScSyg1 and HsXpr1 show a conserved homodimeric structure and the transmembrane part of each protomer consists of 10 TM helices. Helix TM1 establishes the dimer contact by means of a glycine zipper motif, which is a known oligomerization motif. Helices TM2-5 form a hydrophobic pocket that has density for a lipid molecule. Whether the lipid binding into the hydrophobic pocket has an allosteric effect on the phosphate export activity or only serves protein stabilization is not known. Helices TM5-10 form a six-helix bundle, which constitutes a putative phosphate translocation pathway in its center. This bundle is formed by the protein sequence annotated as EXS domain.
The respective phosphate translocation pathways of ScSyg1 and HsXpr1 show structural differences. While the translocation pathway in HsXpr1 is accessible from the cytoplasm, in ScSyg1 it is closed by a large loop of the SPX domain. Interestingly, this loop is not conserved in higher eukaryotes and is therefore not present in HsXpr1. Another difference are distinct conformations of helix TM9. In ScSyg1, TM9 adopts a kinked conformation, which results in the translocation pathway being open to the extracellular side. In contrast, TM9 adopts a straight conformation in HsXpr1, resulting in the placement of a highly conserved tryptophane residue in the middle of the translocation pathway. As a result, the translocation pathway in HsXpr1 is closed to the extracellular side.
The e+e−→D+sDs1(2536)− and e+e−→D+sD∗s2(2573)− processes are studied using data samples collected with the BESIII detector at center-of-mass energies from 4.530 to 4.946~GeV. The absolute branching fractions of Ds1(2536)−→D¯∗0K− and D∗s2(2573)−→D¯0K− are measured for the first time to be (35.9±4.8±3.5)% and (37.4±3.1±4.6)%, respectively. The measurements are in tension with predictions based on the assumption that the Ds1(2536) and D∗s2(2573) are dominated by a bare cs¯ component. The e+e−→D+sDs1(2536)− and e+e−→D+sD∗s2(2573)− cross sections are measured, and a resonant structure at around 4.6~GeV with a width of 50~MeV is observed for the first time with a statistical significance of 15σ in the e+e−→D+sD∗s2(2573)− process. It could be the Y(4626) found by the Belle collaboration in the D+sDs1(2536)− final state, since they have similar masses and widths. There is also evidence for a structure at around 4.75~GeV in both processes.
Using a sample of 448.1×106 ψ(2S) events collected with the BESIII detector, we perform a study of the decay J/ψ→K+K− via ψ(2S)→π+π−J/ψ.
The branching fraction of J/ψ→K+K− is determined to be BK+K−=(3.072±0.023(stat.)±0.050(syst.))×10−4, which is consistent with previous measurements but with significantly improved precision.
In dieser Dissertation werden die Erfahrungen mit verschiedenen Präparationsmethoden für CH-Kavitäten beschrieben, um die Leistung der Kavitäten nach der Herstellung weiter zu steigern. Die Leistung wird anhand von zwei wichtigen HF-Parametern bewertet:
dem elektrischen Feld Ea und der intrinsischen Güte Q0. Im Gegensatz zu normalleitenden (NC) Kavitäten kann die intrinsische Güte von supraleitenden (SC) Kavitäten mit zunehmendem elektrischem Feld erheblich variieren. Das optimale Ergebnis für die Kavitätenpräparation ist die Erhöhung des maximalen elektrischen Feldes unter Beibehaltung eines höheren Q0 über die gesamte Feldspanne. Da Q0 umgekehrt proportional zu den Kavitätsverlusten ist, reduziert eine Erhöhung des Qualitätsfaktors die Kryoverluste für den Betrieb bei gegebenem Feldniveau. Die Entwicklung der Kavitätenperformanz im Verlauf dieser Arbeit dargestellt.
Die meisten SC-Kavitäten sind elliptische Strukturen, welche bei hoher Geschwindigkeit und Tastrate angewendet werden. Die Präparationsmethoden wurden daher überwiegend auf diese Strukturen angewandt und optimiert. Diese Arbeit konzentriert sich auf die Umsetzung der zuverlässigsten und vielversprechendsten Oberflächenbehandlungen mittels des ersten vom IAP entwickelten SC 360MHz CH-Prototyps. Diese Kavität wies nach 11 Jahren Lagerung eine verminderte Leistung auf, welche mit Röntgenstrahlung bei bereits niedrigen elektrischen Feldern einherging. Dies deutet auf eine unbeabsichtigte Belüftung mit normaler Luft hin, durch die Partikel eingeführt wurden, die als verstärkende Quellen von Elektronen fungierten. Außerdem musste der Leistungskoppler aufgrund einer starken Überkopplung neu ausgelegt werden.
Die Kavität wurde für 48 Stunden bei 120◦ C mittels Heizbändern in der Experimentierhalle des IAP’s ausgeheizt, was zu einer Verbesserung des Qualitätsfaktors bei niedrigen Werten und zu einer Verkürzung der für die Konditionierung von Multipacting-Barrieren erforderlichen Zeit führte. Allerdings wurde durch diese Behandlung das maximale erreichbare elektrische Feld weiter verringert. Die Verbesserung der Güte ist auf das Ausgasen der Kohlenwasserstoffe während des Backvorgangs zurückzuführen. Die negative Auswirkung auf das maximale elektrische Feld ist weniger auf das Backen selbst zurückzuführen als auf den Transport der Kavität und die verwendeten Vakuumkomponenten, die in der Versuchshalle gelagert sind.
Die beobachtete Leistungseinschränkung lässt sich hauptsächlich durch Partikel im Inneren des Resonators erklären, da Feldemission bei niedrigen Feldstärken auftrat. Eine Hochdruckspülung mit ultrareinem Wasser (HPR) ist das Standardverfahren, um nach Behandlungen, bei denen das Risiko einer Oberflächenkontamination besteht, eine hohe Reinheit der inneren Oberflächen zu erreichen. Die HPR wurde in Zusammenarbeit mit dem Helmholtz-Institut-Mainz und der Gesellschaft für Schwerionenforschung geplant und durchgeführt. Der Resonator zeigte bereits während der Messung der Q-E-Kurve eine Zunahme der transmittierten Leistung bei konstanter Vorwärtsleistung, was vor der HPR nicht der Fall war. Bei der CW-HF-Konditionierung zeigte die Kavität den höchsten Gradienten bei einem deutlich schwächeren Q-Abfall bei hohen Feldstärken.
Sowohl bei der Messung von 2008 als auch bei der beschriebenen Messung wurde die Kavität mit einer HPR-Behandlung fertiggestellt, aber für die HPR-Behandlung bei HIM in Mainz wurden einige Anpassungen vorbereitet. Der CH Prototyp verfügt über keine zusätzlichen Spülports und wurde daher mit zwei verschiedenen Düsen mit unterschiedlichen Sprühwinkeln gespült, um die erreichbaren inneren Resonatorflächen zu maximieren. Die Verwendung mehrerer Sprühwinkel könnte auch für CH-Kavitäten mit Spülöffnungen von Vorteil sein und sollte für zukünftige HPR-Anwendungen in Betracht gezogen werden.
Die Heliumbehandlung wurde am CH-Prototyp 2,5 Stunden lang durchgeführt und lieferte vielversprechende Ergebnisse in Bezug auf die Güte und die Gradientenoptimierung.
Während dieses Prozesses wurde die emittierte Röntgenstrahlung in Richtung am Arbeitsplatzs gemessen und zeigte starke zeitabhängige Fluktuationen. Dies deutete auf die Beseitigung von Partikeln hin und wurde anschließend durch einen Anstieg des elektrischen Feldes von 8,4 auf 8,7 MV/m bestätigt. Eine unerwartete Auswirkung wurde bei der Q-Steigung im mittleren bis hohen Feld festgestellt, bei der der Qualitätsfaktor im Vergleich zum HF-konditionierten Fall eine Erhöhung von 5% oberhalb von 2MV/m aufwies. Dieser systematische Anstieg wurde für diesen Beschleuniger vor der Behandlung bisher nicht beobachtet. Stickstoffgedopte Kavitäten zeigen ein ähnliches Verhalten, bei dem Wechselwirkungen innerhalb der Oxidschicht mit Änderungen der Qualitätsfaktoren korreliert sind. Da Helium ein nicht reaktives Element ist, sind mögliche Erklärungen für diesen Effekt der Sputterprozess und die Einlagerung von Helium innerhalb der Oberfläche. Eine Serie von Heliumbehandlungen ist geplant, um ein optimiertes und sicheres Rezept für CH-Kavitäten zu finden. Die Q-E-Messung nach der Abkühlung und vor der Behandlung wird auch zeigen, ob der Leistungsgewinn durch ein Aufwärmen auf Raumtemperatur beeinträchtigt wird.
Die in dieser Arbeit skizzierte Behandlungssequenz wird für CH-Kavitäten dringend empfohlen. Das Ausheizen hat sich bei der Verringerung des Multipactings and der Güteabnahme bei hohen Feldern als wirksam erwiesen und bleibt von der anschließenden HPR unbeeinflusst. In dieser Arbeit wurden keine negativen Auswirkungen der HPR auf das Multipactingverhalten festgestellt. Anschließend wird eine CW-HF-Konditionierung durchgeführt, bis keine weitere Leistungszunahme der Kavität mehr zu verzeichnen ist.
Wenn die Kavität immer noch durch Feldemission begrenzt ist, sollte eine Wiederholung der HPR-Behandlung in Betracht gezogen werden, da bei sorgfältiger Durchführung der HPR keine der bisherig gefertigten CH-Kavitäten hierdurch begrenzt war. Es ist auch anzumerken, dass die Heliumbehandlung nur an der 360MHz CH-Kavität durchgeführt wurde, als diese eine geringe Strahlung durch Feldemission aufwies. Das Risiko des Heliumprocessing an CH-Kavitäten unter starker Feldemission ist unbekannt. Es ist zu erwarten, dass die Elektronenströme und damit die Ionenbeschusslawinen zunehmen und ein größeres Risiko für die Beschädigung von der Komponenten darstellen. Nach dem derzeitigen Kenntnisstand sollte die Heliumbehandlung nur für gut vorbereitete Kavitäten mit minimaler Feldemission in Betracht gezogen werden.
An independent Taiwanese lineage of powdery mildew on the endemic host species Koelreuteria henryi

Background: Powdery mildews (Erysiphaceae, Ascomycota) are common plant disease agents and also cause stress for forest and fruit trees worldwide as well as in Taiwan. The powdery mildew Erysiphe bulbouncinula on Koelreuteria host trees was considered an endemic species in China. While in China the host was K. paniculata and only the teleomorph stage found, the anamorph and the teleomorph were both recorded for the host in Taiwan, K. henryi. We aimed to clarify the relationship of the powdery mildews recorded under E. bulbouncinula with an apparently disjunct distribution.
Results: Specimens of powdery mildew on K. henryi from Taiwan were characterized based on the anamorph morphology and DNA sequences. They revealed a new record of Sawadaea koelreuteriae for this host species and Taiwan and a new species of Erysiphe, E. formosana, sister to E. bulbouncinula from China.
Conclusions: In Erysiphe on Koelreuteria hosts, speciation of plant parasitic fungi seems to be correlated with disjunct host and geographic distribution possibly shaped by extinction of potential host species which are known only as fossils. Two of the three extant East Asian species of Koelreuteria are now known as hosts of specific Erysiphe species. We may predict a further not yet discovered Erysiphe species on the third East Asian species, K. bipinnata, in South and Southwest China. In the speciation in Sawadaea, the extinction events in Koelreuteria can be excluded from being involved.
Das partizipative Forschungsprojekt "Allein aber vernetzt? Digitale (Un)gleichheiten und soziale Netzwerke bei alleinlebenden Menschen" beschäftigte sich von 2021-2024 mit Alleinlebenden im Alter, ihren Beziehungsnetzwerken und der Frage welche Rolle digitale Geräte und die Digitalisierung aller Lebensbereiche in diesem Zusammenhang spielen. Durch die Zusammenarbeit mit einer Gruppe von Ko-Forschenden in Frankfurt und Egelsbach stand die Perspektive der Gruppe im Mittelpunkt.
Im vorliegenden White Paper wird das Projekt sowie zentrale Ergebnisse und die erarbeiteten Handlungsempfehlungen der deutschen Teilstudie vorgestellt. Diese wurde vom BMBF gefördert und ist Teil des europäischen Verbundprojektes "EQualCare - Alone but connected? Digital (in)equalities in care work and generational relationships among older people living alone" der Joint Programming Initiative (JPI) “More Years Better Lives”.

Based on 4.5 fb−1 of e+e− collision data accumulated at center-of-mass energies between 4599.53 MeV and 4698.82 MeV with the BESIII detector, the decay Λ+c→nK0Sπ+π0 is observed for the first time with a significance of 9.2σ. The branching fraction is measured to be (0.85±0.13±0.03)%, where the first uncertainty is statistical and the second systematic, which differs from the theoretical prediction based on isospin by 4.4σ. This indicates that there may be resonant contributions or some unknown dynamics in this decay.
We report results on an elastic cross section measurement in proton-proton collisions at a center-of-mass energy s√=510 GeV, obtained with the Roman Pot setup of the STAR experiment at the Relativistic Heavy Ion Collider (RHIC). The elastic differential cross section is measured in the four-momentum transfer squared range 0.23≤−t≤0.67 GeV2. We find that a constant slope B does not fit the data in the aforementioned t range, and we obtain a much better fit using a second-order polynomial for B(t). The t dependence of B is determined using six subintervals of t in the STAR measured t range, and is in good agreement with the phenomenological models. The measured elastic differential cross section dσ/dt agrees well with the results obtained at s√=546 GeV for proton--antiproton collisions by the UA4 experiment. We also determine that the integrated elastic cross section within the STAR t-range is σfidel=462.1±0.9(stat.)±1.1(syst.)±11.6(scale) μb.
Vor dem Hintergrund der epochalen Verunsicherungen des ausgehenden 18. Jahrhunderts stellen sich Goethe und Schiller in ihrer Dramatik den politischen Dimensionen und Abgründen der klassischen französischen Tragödie. Verkappte Staatsgründungen beherrschen ebenso das Geschehen wie der Staatsstreich, der Bürgerkrieg oder der failing state. Eine neue Aktualität gewann das Drama Corneilles und Racines jedoch erst wegen seiner Aufladung der klassischen Form mit staatspolitischen Fundamentalfragen. Das Ringen um eine Regelpoetik und ihre angemessene Umsetzung, vermeintliche Nebensächlichkeiten wie die Einheit der Zeit werden zum Schauplatz genuin politischer Reflexion. Goethe und Schiller verzichten auf eine Restauration der klassischen Form, sie entdecken aber ihre ursprüngliche Energie wieder und verhandeln diese neu. Besonders deutlich zeigt sich dies am dramaturgischen Konflikt zwischen König und Held, der einen bislang weithin übersehenen Grundpfeiler der klassischen Tragödie bildet.
The ALICE experiment at the LHC investigates the properties of the hot and dense nuclear matter created in heavy-ion collisions. By comparing the particle production in pp and p-Pb collisions, possible nuclear initial state effects can be isolated. Measurements of the ω meson pT-spectra in pp and p-Pb collisions not only allow for a determination of the nuclear modification factor RpPb, but also provide insight into the fragmentation process and serve as vital input for decay background simulations for direct photons. In this contribution, measurements of the ω meson production in pp and p-Pb collisions at √sNN=5.02 TeV are presented. This includes the signal extraction and various corrections of the ω meson yields, leading to their production cross sections and the first measured nuclear modification factor RpPb of the ω meson at LHC energies.

• Solidification and cooling of an intruded dyke or sill within the middle or shallow crust generate stresses of order 200 MPa, which relax on time scales of a few years to million years.
• Stresses may exceed the brittle strength forming tensile fractures.
• Combined with the pressure gradient within the over-pressurized felsic melts, this explains the migration of felsic contact melt into shrinkage cracks (Sederholm-type veins).
Abstract
Rapid emplacement of a mafic dyke or sill at mid-crustal depth heats and possibly melts the felsic wall rock followed by solidification. Associated volume changes generate stresses, possibly enforcing brittle failure and melt migration. We model the evolution of melting, solidification, temperature, and stress including visco-elastic relaxation in 1D - dykes or -sills using realistic rock rheologies of the Weschnitz pluton (Odenwald). For deep emplacement (Case 1, 15.3 km) extensive contact melting of the wall rock occurs, for shallow emplacement (Case 2, 10 km) it is negligible. The stresses are zero at high melt fractions, but increase during solidification and cooling: The intrusion orthogonal stress is always zero. The intrusion parallel stress σ‖ within the intrusion is tensile (O(200 MPa)). It relaxes on a time scale between a few years (Case 1) and 0.6 m.y. (Case 2). Within the wall rock σ‖ is compressive during heating, but becomes tensile under solidification and cooling. Wall rock stresses relax on a time scale of months to 100 years. A Deborah number is defined based on viscous to thermal relaxation allowing generalization of our results. Adding lithostatic stresses, the total stresses of Case 1 remain below the brittle strength, while for Case 2 they may exceed it. Adding the lithostatic pressure to the melt pressure, the effective stresses exceed the brittle strength and intrusion orthogonal tensile fractures are predicted. Combined with the pressure gradient within the over-pressurized felsic melts generated in the wall rock, this explains the migration of felsic contact melt into shrinkage cracks of the mafic sill in the Weschnitz pluton.
Der Beitrag thematisiert die literarische Darstellung virtueller Investigationen, welche eine performativ-schauspielerische Wiederholung von Tatgeschehnissen beinhalten. Er analysiert insbesondere die Rolle von Zufallsmotiven und -aspekten in solchen Darstellungen und deren metapoetische Bedeutung im Rahmen einer Gattungspoetik des Kriminalromans. Als Fallbeispiele dienen hierfür Friedrich Dürrenmatts 'Requiem auf den Kriminalroman' "Das Versprechen" und Stanisław Lems Roman "Der Schnupfen". Im Vergleich der beiden Texte zeigt sich unter anderem, dass die Idee eines genauen 're-enactment' von Tathergängen auf der Grundlage virtueller Rekonstruktionen in beiden Romanplots nicht direkt zum gewünschten Ergebnis führt, dass aber in Lems Roman eine Integration des Faktors Zufall in virtuelle Ermittlungsprozesse und in die Poetik des Kriminalromans insgesamt sehr viel positiver gewertet wird, während der Einfluss des Zufalls bei Dürrenmatt ebenfalls als unhintergehbar, aber nicht als uneingeschränkt begrüßenswert perspektiviert wird.
Natural Language Processing (NLP) for big data requires an efficient and sophisticated infrastructure to complete tasks both fast and correctly. Providing an intuitive and lightweight interaction with a framework that abstracts and simplifies complex tasks assists in reaching this goal. This bachelor thesis extends the NLP framework Docker Unified UIMA Interface (DUUI) by an API and a web-based graphical user interface to control and manage pipelines for automated analysis of large quantities of natural language. The extension aims to reduce the entry barrier into the field as well as to accelerate the creation and management of pipelines according to UIMA standards. Pipelines can be executed in the browser or using the web API directly and then monitored on a document level. The evaluation in usability and user experience indicates that the implementation benefits the framework by making its usage more user friendly, lightweight, and intuitive while also making the management of pipelines more efficient.
The differential cross section for 𝑍0 production, measured as a function of the boson’s transverse momentum (𝑝T), provides important constraints on the evolution of the transverse momentum dependent parton distribution functions (TMDs). The transverse single spin asymmetry (TSSA) of the 𝑍0 is sensitive to one of the polarized TMDs, the Sivers function, which is predicted to have the opposite sign in 𝑝 + 𝑝 → 𝑊 ∕𝑍 + 𝑋 from that which enters in semi-inclusive deep inelastic scattering. In this Letter, the STAR Collaboration reports the first measurement of the 𝑍0∕𝛾∗ differential cross section as a function of its 𝑝T in 𝑝+𝑝 collisions at a center-of-mass energy of 510 GeV, together with the 𝑍0∕𝛾∗ total cross section. We also report the measurement of 𝑍0∕𝛾∗ TSSA in transversely polarized 𝑝+𝑝 collisions at 510 GeV.
The dynamics of the torsion field is analyzed in the framework of the Covariant Canonical Gauge Theory of Gravity (CCGG), a De Donder–Weyl Hamiltonian formulation of gauge gravity. The action is quadratic in both, the torsion and the Riemann–Cartan tensor. Since the latter adds the derivative of torsion to the equations of motion, torsion is no longer identical to spin density, as in the Einstein–Cartan theory, but an additional propagating degree of freedom. As torsion turns out to be totally anti-symmetric, it can be parametrised via a single axial vector. It is shown in this paper that, in the weak torsion limit, the axial vector obeys a wave equation with an effective mass term which is partially dependent on the scalar curvature. The source of torsion is thereby given by the fermion axial current which is the net fermionic spin density of the system. Possible measurable effects and approaches to experimental analysis are addressed. For example, neutron star mergers could act as a dipoles or quadrupoles for torsional radiation, and an analysis of radiation of pulsars could lead to a detection of torsion wave background radiation.
Autophagy is an important degradation pathway mediating the engulfment of cellular material (cargo) into autophagosomes followed by degradation in autophagosomes.
Different stress stimuli, e.g. nutrient deprivation, oxidative stress or organelle damage, engage autophagy to maintain cellular homeostasis, recycle nutrients or remove damaged cell organelles. Autophagy not only degrades bulk cytoplasmic material but also selective autophagic cargo, for example lysosomes (lysophagy), mitochondria (mitophagy), ER (ER-phagy), lipid droplets (lipophagy), protein aggregates (aggrephagy) or pathogens (xenophagy). Selective autophagy pathways are regulated by selective autophagy receptors which bind to ubiquitinated cargo proteins and link them to LC3 on the autophagosomal membrane.
Ubiquitination is an essential post-translational modification controlling different cellular processes such as proteasomal and lysosomal degradation or innate immune signaling.
M1-linked (linear) poly-Ubiquitin (poly-Ub) chains are exclusively assembled by the E3 ligase linear ubiquitin chain assembly complex (LUBAC) and removed by the M1 poly-Ub-specific OTU domain-containing deubiquitinase with linear linkage specificity (OTULIN). In addition to key functions in innate immune signaling and nuclear factor-κB (NF-κB) activation, M1 ubiquitination is also implicated in the regulation of autophagy.
LUBAC and OTULIN control autophagy initiation and maturation and the autophagic clearance of invading bacteria via xenophagy. However, additional functions of LUBAC- and OTULIN-regulated M1 ubiquitination in autophagy are largely unknown and it also remains unexplored if LUBAC and OTULIN control other selective autophagy pathways in addition to xenophagy. This study aimed to unravel the role of LUBAC- and OTULIN-controlled M1 ubiquitination in bulk and selective autophagy in more detail.
In this study, characterization of OTULIN-depleted MZ-54 glioblastoma (GBM) cells revealed that OTULIN deficiency results in enhanced LC3 lipidation in response to autophagy induction and upon blockade of late stage autophagy with Bafilomycin A1 (BafA1). Furthermore, electron microscopy analysis showed that OTULIN-deficient cells have an increased number of degradative compartments (DGCs), confirming enhanced autophagy activity upon loss of OTULIN. APEX2-based autophagosome content profiling identified various OTULIN-dependent autophagy cargo proteins. Among these were the autophagy receptor TAX1BP1 which regulates different forms of selective autophagy (e.g. lysophagy, aggrephagy) and the glycan-binding protein galectin-3 which serves key functions in lysophagy, suggesting a role of OTULIN and M1 poly-Ub in the regulation of aggrephagy and lysophagy.

To study aggrephagy, protein aggregation was induced with puromycin which causes premature termination of translation and accumulation of defective ribosomal products (DRiPs). Loss of OTULIN increased the number of M1 poly-Ub-positive foci and insoluble proteins and reduced the levels of soluble TAX1BP1 and p62 in response to puromycin-induced proteotoxic stress.
Intriguingly, upon induction of lysosomal membrane permeabilization (LMP) with the lysosomotropic drug L-Leucyl-L-Leucine methyl ester (LLOMe), M1 poly-Ub strongly accumulated at damaged lysosomes and colocalized with TAX1BP1- and galectin-3-positive puncta. M1 poly-Ub-modified lysosomes formed a platform for NF-κB essential modulator (NEMO) and inhibitor of κB (IκB) kinase (IKK) complex recruitment and local NF-κB activation in a K63 poly-Ub- and OTULIN-dependent manner. Furthermore, inhibition of lysosomal degradation enhanced LLOMe-induced cell death, suggesting pro-survival functions of lysophagy following LMP. Enrichment of M1 poly-Ub at damaged lysosomes was also observed in human dopaminergic neurons and in primary mouse embryonic cortical neurons, confirming the importance of M1 poly-Ub in the response to lysosomal damage.
Together, these results identify OTULIN as a negative regulator of autophagy induction and the autophagic flux and reveal OTULIN-dependent autophagy cargo proteins.
Furthermore, this study uncovers novel and important roles of M1 poly-Ub in the response to lysosomal damage and local NF-κB activation at damaged lysosomes.

Abstract
We study the impact of immigrant legalization on fiscal transfers from state to local governments in the United States, exploiting variation in legal status from the 1986 Immigration Reform and Control Act (IRCA). State governments allocate more resources to IRCA counties, an allocation that is responsive to the electoral incentives of the governor. Importantly, the effect emerges prior to the enfranchisement of the IRCA migrants and we argue it is driven by the IRCA’s capacity to politically empower already legal Hispanic migrants in mixed legal status communities. The IRCA increases turnout in large Hispanic communities as well as Hispanic political engagement, without detectably triggering anti-migrant sentiment.
Although, during the past decades, substantial advances emerged in identifying major local and systemic factors contributing to initiation and progression of osteoarthritis (OA), some neuroendocrine mechanisms are still not understood or even neglected when thinking about novel therapeutic options. One of which is the sympathetic nervous system that exhibits various OA-promoting effects in different tissues of the joint. Interestingly, the β2-adrenoceptor (AR) mediates the majority of these effects as demonstrated by several in vitro, in vivo as well as in clinical studies. This review article does not only summarize studies of the past two decades demonstrating that the β2-AR plays an OA-promoting role in different tissues of the joint but also aims to encourage the reader to think about next-level research to discover novel and innovative preventive and/or therapeutic strategies targeting the β2-AR in OA.
We report a new measurement of transverse single-spin asymmetries for dijet production in collisions of polarized protons at s√ = 200 GeV. Correlations between the proton spin and the transverse momenta of its partons, each perpendicular to the proton momentum direction, are probed at high Q2 ≈160 GeV2. The associated Sivers observable ⟨kT⟩, the average parton transverse momentum, is extracted using simple kinematics. Nonzero Sivers effects are observed for the first time in dijets from proton-proton collisions, but only when the jets are sorted by their net charge, which enhances the u- or d-quark contributions to separate data samples. This also enables a simple kinematic approach for determination of the individual partonic contributions to the observed asymmetries.
We report the first measurements of cumulants, up to 4𝑡ℎ order, of deuteron number distributions and protondeuteron correlations in Au+Au collisions recorded by the STAR experiment in phase-I of Beam Energy Scan (BES) program at the Relativistic Heavy Ion Collider. Deuteron cumulants, their ratios, and proton-deuteron mixed cumulants are presented for different collision centralities covering a range of center-of-mass energy per nucleon pair √𝑠NN = 7.7 to 200 GeV. It is found that the cumulant ratios at lower collision energies favor a canonical ensemble over a grand canonical ensemble in thermal models. An anti-correlation between proton and deuteron multiplicity is observed across all collision energies and centralities, consistent with the expectation from global baryon number conservation. The UrQMD model coupled with a phase-space coalescence mechanism qualitatively reproduces the collision-energy dependence of cumulant ratios and proton-deuteron correlations.

Dynamic imaging of landmark organelles, such as nuclei, cell membrane, nuclear envelope, and lipid droplets enables image-based phenotyping of functional states of cells. Multispectral fluorescent imaging of landmark organelles requires labor-intensive labeling, limits throughput, and compromises cell health. Virtual staining of label-free images with deep neural networks is an emerging solution for this problem. Multiplexed imaging of cellular landmarks from scattered light and subsequent demultiplexing with virtual staining saves the light spectrum for imaging additional molecular reporters, photomanipulation, or other tasks. Published approaches for virtual staining of landmark organelles are fragile in the presence of nuisance variations in imaging, culture conditions, and cell types. This paper reports model training protocols for virtual staining of nuclei and membranes robust to label-free imaging parameters, cell states, and cell types. We developed a flexible and scalable convolutional architecture, named UNeXt2, for supervised training and self-supervised pre-training. The strategies we report here enable robust virtual staining of nuclei and cell membranes in multiple cell types, including neuromasts of zebrafish, across a range of imaging conditions. We assess the models by comparing the intensity, segmentations, and application-specific measurements obtained from virtually stained and experimentally stained nuclei and membranes. The models rescue the missing label, non-uniform expression of labels, and photobleaching. We share three pre-trained models, named VSCyto3D, VSCyto2D, and VSNeuromast, as well as VisCy, a PyTorch-based pipeline for training, inference, and deployment that leverages the modern OME-Zarr format.
Cyclin CLB2 mRNA localization and protein synthesis link cell cycle progression to bud growth

Clb2 is a conserved mitotic B-type cyclin, the levels of which are finely controlled to drive progression through the cell cycle. While it is known that CLB2 transcription and Clb2 protein degradation are important for precise control of its expression, it remains unclear whether the synthesis of Clb2 is also regulated. To address whether and how Clb2 expression levels respond to cell growth changes and adapt cell cycle progression, we combined single-cell and single-molecule imaging methods to measure CLB2 mRNA and protein expression throughout the Saccharomyces cerevisiae cell cycle. We found that the CLB2 mRNA was efficiently localized to the yeast bud as soon as this compartment was formed, but strikingly the Clb2 protein accumulated in the mother nucleus. The CLB2 mRNA localization in the yeast bud by the She2-3 complex did not control protein localization but rather promoted CLB2 translation. Moreover, CLB2 mRNA bud localization and protein synthesis were coupled and dependent on a single secondary structure -a ZIP code-located in the coding sequence. In a CLB2 ZIP code mutant, mRNA localization was impaired and Clb2 protein synthesis decreased, resulting in changes in cell cycle distribution and increased size of daughter cells at birth. Finally, while in WT cells the Clb2 protein concentration followed bud growth, this relationship was impaired in the ZIP code mutant. We propose that S. cerevisiae couples the control of CLB2 mRNA bud localization and protein synthesis to coordinate cell growth and cell cycle progression. This mechanism extends our knowledge of CLB2 expression regulation, and constitutes a novel function for mRNA localization.
Im Rahmen dieser Doktorarbeit werden drei Schwerpunkte behandelt: 1) Die hocheffektive Beschleunigung von Elektronen und Protonen durch die Wechselwirkung von relativistischen Laserpulsen mit Schäumen. 2) Die Erzeugung und Messung hochintensiver Betatronstrahlung von direkt laserbeschleunigten (DLA-) Elektronen. 3) Die Anwendung von DLA-Elektronen für den biologischen FLASH-Effekt mit einer rekordbrechenden Dosisrate.
Die direkte Laserbeschleunigung von Elektronen wurde durch die Wechselwirkung eines sub-ps-Laserpulses mit einer Intensität von ~ 10^19 W/cm^2 mit einem Plasma nahe kritischer Elektronendichte (NCD) untersucht. Ein sub-mm langes NCD-Plasma wurde durch Erhitzen eines Schaums mit einer niedrigen Dichte mit einem ns-Puls von 10^13-10^14 W/cm^2 erzeugt. Die Experimente wurden an der PHELIX-Anlage (Petawatt Hoch- Energie Laser für Schwerionenexperimente) in den Jahren 2019 – 2023 durchgeführt. Während der Suche nach optimalen Bedingungen für die Beschleunigung von Elektronen und Protonen wurden die Parameter des ns-Pulses variiert und verschiedene Targets verwendet. Es wurde gezeigt, dass das Plasma im Schaum gute Voraussetzungen für die Erzeugung gerichteter, ultrarelativistischer DLA-Elektronen mit Energien von bis zu 100 MeV bietet. Die Elektronen weisen eine Boltzmann-ähnliche Energieverteilung mit einer Temperatur von 10-20 MeV auf.
Optimale Bedingungen für eine effektive Beschleunigung von DLA-Elektronen wurden bei der Kombination eines CHO-Schaums mit einer Dichte von 2 mg/cm3 und einer Dicke von 300-500 µm mit einer Metallfolie erreicht. Die Gesamtladung der detektierten Elektronen mit Energien über 1,5 MeV erreichte 0,5-1 µC mit der Umwandlungseffizienz der Laserenergie von ~ 20-30%.
Außerdem wird die Beschleunigung von Protonen durch DLA-Elektronen anders verursacht als bei typischer Target Normal Sheath Acceleration (TNSA). Für die Untersuchung der lokalen Protonenenergieverteilung wurden Magnetspektrometer unter verschiedenen Winkeln zur Laserachse verwendet. Dafür wurde eine Filtermethode entwickelt, welche es ermöglicht, Spektren von Protonen mit Energien von bis zu 100 MeV zu rekonstruieren. Es wurde gezeigt, dass am PHELIX durch die Kombination von einem ~ 300-400 µm dicken CHO-Schaum mit einer Dichte von 2 mg/cm^3 und einer 10 µm dicken Au-Folie bei einer Intensität des sub-ps-Pulses von ~ 10^19 W/cm^2 und unter Verwendung eines optimierten ns-Vorpulses eine optimale Protonenbeschleunigung erreicht wurde. Es wurde ein TNSA-ähnliches Regime mit einer maximalen Cut-off-Energie von 34±0,5 MeV beobachtet. Im Vergleich dazu wurde bei der typischen TNSA unter Verwendung einer 10 µm dicken Au-Folie als Target und derselben Laserintensität eine maximale Cut-off-Energie von 24±0,5 MeV gemessen. Darüber hinaus beobachteten wir einen sehr schwachen Abfall der Protonenanzahl in Abhängigkeit von der Protonenenergie (anders als bei der typischen TNSA) und eine sehr regelmäßige Protonenstrahlverteilung in einem breiten Winkelbereich bis zu hohen Energien. Dies könnte zur Verbesserung der Qualität der Protonenradiographie von Plasmafeldern genutzt werden.
Beim DLA-Prozess (im NCD-Plasma) entsteht Betatronstrahlung durch die Oszillationen von Elektronen in quasi-statischen elektrischen und magnetischen Feldern des Plasmakanals. Um diese Strahlung zu untersuchen, wurde ein neues modifiziertes Magnetspektrometer (X-MS) konstruiert. Das X-MS ermöglicht die 1D-Auflösung mehrerer Quellen. Dank dieser Spezifikation war es möglich, Betatronstrahlung von Bremsstrahlung der ponderomotorischen Elektronen im Metallhalter zu trennen und zu messen.
Im Experiment mit einem CHO-Schaum mit einer Dichte von 2 mg/cm^3 und einer Dicke von ~ 800 µm als Target wurde die von den optimierten DLA-Elektronen erzeugte Betatronstrahlung gemessen. Bei einer Peak-Intensität des dreieckigen ns-Pulses von ~ 3·10^13 W/cm^2 und des sub-ps-Pulses von ~ 10^19 W/cm^2, welcher 4±0,5 ns gegenüber dem ns-Puls verzögert war, betrug der Halbwinkel im FWHM-Bereich des Elektronenstrahls 17±2°. Unter diesen Bedingungen war die Betatronstrahlung mit einem Halbwinkel im FWHM-Bereich von 11±2° für die Photonen mit Energien über 10 keV ebenfalls gerichtet. Die Photonenanzahl mit Energien über 10 keV wurde auf etwa 3·10^10 / 3·10^11 (gerichtete Photonen / Photonen im Halbraum entlang der Laserstrahlrichtung) abgeschätzt. Die maximale Photonenanzahl pro Raumwinkel betrug ~2·10^11 photons/sr. Die Brillanz der registrierten Betatronstrahlung erreichte ~ 2·10^20 photons/s/mm^2/mrad^2/(0.1% BW) bei 10 keV.
Die Verwendung eines Hochstromstrahls aus DLA-Elektronen für die FLASH-Strahlentherapie ermöglicht das Erreichen einer Dosis von bis zu 50-70 Gy während eines sub-ps-Laserpulses. Im Jahr 2021, während der P213-Strahlzeit am PHELIX wurde der Sauerstoffkonzentrationsabfall bei der Bestrahlung von Medien (Wasser und andere biologische Medien) mit DLA-Elektronen in Abhängigkeit von der Dosis untersucht. Die Strahlendosis wurde hierbei indirekt gemessen. Hierfür wurde eine Rekonstruktionsmethode entwickelt, die es ermöglicht, die Dosis innerhalb des „Wasser-Containers“ auf Basis von Messungen außerhalb des Containers mit einem untersuchten Medium zu ermitteln. Es wurde eine gute Übereinstimmung zwischen dem Experiment und einer Monte-Carlo-Simulation für Wasser gezeigt. Die registrierte Dosisrate erreichte einen Rekordwert von ~ 70 TGy/s.
Carbonate archives record a brief snapshot of the ambient Earth’s surface conditions at their deposition. However, the geologically reasonable extraction and interpretation of geochemical proxy data from ancient, diagenetically altered rock archives is fraught with problems. Three issues stand out: the dichotomy between petrographic and geochemical alteration; the lack of quantitative age constraints for specific diagenetic phases resulting in a poorly constrained admixture of local, basin-wide and over-regional (far-field) features; and an often insufficient understanding of the temperatures and compositions of diagenetic fluids. Here, the archive of Devonian marine limestones exposed to multiple far-field diagenetic events is used as an example to explore the above-listed issues. Methods applied include petrography, micro XRF, fluid inclusion data, clumped isotopes, δ13C and δ18O isotopes, 87Sr/86Sr ratios and quartz trace element data. Devonian limestones studied here were overprinted by two cross-cutting regional fault zones (T ≈ 230 °C) by multiple events between the Variscan Orogeny and the late Paleogene. The following processes are recorded: (i) protolith deposition and partial dolomitisation during rapid burial in the Middle/Late Devonian (T ≈ 180 °C); (ii) deep burial to ca 6.5 km and tectonic/hydrothermal overprint during the Variscan Orogeny in the Carboniferous (T ≈ 90–230 °C); (iii) rapid uplift to 1–2 km burial depth at the end of the Variscan Orogeny and hypogene karstification (T ≈ 50 to 100 °C) initiated by regional geology in the Permian/Triassic; (iv) tectonic/hydrothermal overprint during the opening of the Proto-Atlantic Ocean between the Early Jurassic and the Early Cretaceous (T ≈ 50 to 130 °C); (v) tectonic/hydrothermal overprint including renewed hypogene karstification and hydrothermal calcite cement precipitation (T ≈ 50 to 180 °C) during Alpine Orogeny between the Late Cretaceous and late Paleogene. Despite this complex series of diagenetic events, the protolith limestones largely preserved their respective Middle/Late Devonian dissolved inorganic carbon (DIC) and 87Sr/86Sr signatures. This study documents that geochemical proxy data, placed into their petrographic, paleotemperature, and local to over-regional context, significantly increases the ability to extract quantitative information from ancient carbonate rock archives. Research shown here has wider relevance for carbonate archive research in general.
Das Schilddrüsenkarzinom (SK) ist die häufigste bösartige endokrine Tumorerkrankung. Während das nicht-metastasierte und nicht-mutierte papilläre Schilddrüsenkarzinom (PSK) und das follikuläre Schilddrüsenkarzinom (FSK) eine gute Heilungschance aufweisen, zeigen die mutierten und metastasierten Varianten des PSK und FSK sowie das anaplastische Schilddrüsenkarzinom (ASK) weiterhin eine schlechte Prognose. Die Entwicklung von Therapieresistenzen stellen hierbei ein Hauptproblem in der Behandlung des fortgeschrittenen Schilddrüsenkarzinoms dar.
In den letzten Jahren wurden in Studien zunehmend Tumor-initiierende Zellen (TIZ) beschrieben, welche eine kleine Subpopulation von Zellen mit der Fähigkeit zur Selbsterneuerung, Tumorinitiierung und Entwicklung von Therapieresistenzen von Tumoren darstellen. Die Existenz von TIZ wurde auch im SK nachgewiesen. Ein entscheidender Faktor für die Persistenz von TIZ ist die Hypoxie, welche über eine Veränderung des Tumormikromilieus und des Zellmetabolismus zur Entstehung von Therapieresistenzen beiträgt. Ein durch Hypoxie hochreguliertes Enzym ist die Carboanhydrase IX (CAIX). CAIX wird hauptsächlich von Tumorzellen exprimiert und katalysiert die Reaktion von Kohlendioxid zu Bicarbonat und einem Proton und trägt damit zur Säurepufferung der Tumorzelle bei. CAIX stellt somit einen entscheidenden Faktor für das Überleben von Tumorzellen in einem hypoxischen Milieu dar. Des Weiteren ist eine erhöhte Expression von CAIX mit einem schlechten Patienten-Outcome assoziiert, wie z.B. im Brustkrebs. Diese Eigenschaften machen CAIX zu einem attraktiven Angriffspunkt einer zielgerichteten Tumortherapie. Die vorliegende Studie hat zum Ziel, die Expression von CAIX sowie dessen biologische Rolle im Schilddrüsenkarzinom näher zu untersuchen.
Hierzu wurden Proben von 114 SK-Patienten immunhistochemisch auf eine CAIX-Expression untersucht und mit tumorfreiem Schilddrüsengewebe verglichen. Hierbei waren unterschiedliche SK-Subtypen vertreten. Für eine weitere Validierung der Expressionsdaten erfolgte die Auswertung eines Datasets von „The Cancer Genome Atlas“ (TCGA) mithilfe von cBioportal. Da die Hypoxie ein wichtiger Faktor für die Persistenz von TIZ ist, wurde die CAIX-Expression in Tumorsphären, ein in vitro Nachweis von TIZ-Aktivität, mittels der Durchflusszytometrie bestimmt und mit der CAIX-Expression von Monolayern verglichen. Als SK-Zelllinien wurden BCPAP (PSK), FTC 133 (FSK) und 8505 C (ASK) verwendet. Anschließend wurde mithilfe der Polymerasekettenreaktion und Immunofluoreszenzfärbung untersucht, ob eine CAIX-Expression in den Tumorsphären mit der Expression von bereits bekannten Stammzellmarkern, u.a. NANOG, assoziiert ist. Die Unterschiede der CAIX-Expression, nach Inkubation der Monolayer jeweils in Normoxie und Hypoxie, wurden mittels Durchflusszytometrie bestimmt. Mithilfe eines genetischen CAIX-Knockdowns sowie einer pharmakologischen Inhibition mit dem CAIX-Inhibitor Methazolamid (MZM) wurde die Tumorzellproliferation und -Sphärenbildung unter Normoxie und Hypoxie bestimmt. Zusätzlich wurde der Einfluss von MZM auf die Apoptose und den Zellzyklus untersucht.
Immunhistochemische Färbungen der Gewebeproben von SK-Patienten zeigten, dass die CAIX-Expression sowohl im PSK und FSK als auch im ASK im Vergleich zum tumorfreien Schilddrüsengewebe erhöht war. Des Weiteren zeigte die klinisch-pathologische Datenanalyse, dass eine erhöhte CAIX-Expression mit dem Auftreten von Lymphknotenmetastasen im differenzierten SK assoziiert war. Auch die Analyse des TCGA-Datasets bestätigte, dass eine erhöhte Expression der CAIX-mRNA mit einem fortgeschrittenen Tumorstadium, Fernmetastasen und mit einem kürzeren Gesamt-Überleben von SK-Patienten korrelierte. Die weiteren funktionellen in vitro Untersuchungen ergaben, dass die CAIX-Expression in den Tumorsphären im Vergleich zu Monolayern erhöht und mit einer erhöhten Expression von Stammzellmarkern assoziiert war. Ein genetischer CAIX-Knockdown und eine CAIX-Inhibition mit MZM führten über eine Induktion der Apoptose und eines Zellzyklusarrests zu einer verminderten Tumorzellproliferation und Sphärenbildung.
Zusammenfassend deuten die Ergebnisse darauf hin, dass CAIX ein vielversprechendes Zielmolekül für eine gezielte Tumortherapie des fortgeschrittenen SK ist. Um diese Hypothese bestätigen zu können, sind jedoch weitere prospektive Analysen von Patientenproben sowie funktionelle in vivo Untersuchungen am SK nötig.

Abstract
We conducted scaled analogue modelling to show the influence of varying single layer initial orientation on the geometry of folds and boudins in a bulk constrictional strain field. The initial angle between the plane of shortening and the competent layer (θZ(i)) was incrementally increased from 0° to 90° by multiples of 11.25°. While the amount of layer thickening decreased with increasing θZ(i), the deformation structures produced range from pure dome-and-basin folds to coeval folds and boudins. Based on the attitude of fold axes, there are extension-parallel (FEPR) and extension-perpendicular (FEPP) folds, with axes subparallel and subperpendicular to the principal stretching axis (X), respectively. Coeval growth of FEPR folds and boudins occurred when θZ(i) > ca. 25°. The FEPP folds can be subdivided into a first type which affect the entire layer (if θZ(i) ranges between 11.25 and 78.75°) and a second type, referred to as FBEPP folds, which are affecting pre-existing boudins if θZ(i) > 45°. The interlimb angle of all types of folds increases with increasing θZ(i). Folds and boudins similar to the ones produced in this study can be found in salt domes and in tectonites of subduction zones.
Rationale: Potassium (K) is a major component of several silicate minerals and seawater, and, therefore, constraining past changes in the potassium cycle is a promising way of tracing large-scale geological processes on Earth. However, [K] measurement using inductively coupled plasma mass spectrometry (ICP-MS) is challenging due to an ArH+ interference, which may be of a similar magnitude to the K+ ion beam in samples with <0.1% m/m [K].
Methods: In this work, we investigated the effect of the ArH+ interference on K/Ca data quality by comparing results from laser-ablation (LA)-ICP-MS measured in medium and high mass resolution modes and validating our LA results via solution ICP-optical emission spectroscopy (OES) and solution ICP-MS measurements. To do so, we used a wide range of geological reference materials, with a particular focus on marine carbonates, which are potential archives of past changes in the K cycle but are typically characterised by [K] < 200 μg/g. In addition, we examine the degree to which trace-element data quality is driven by downhole fractionation during LA-ICP-MS measurements.
Results: Our results show that medium mass resolution (MR) mode is sufficiently capable of minimising the effect of the ArH+ interference on K+. However, the rate of downhole fractionation for Na and K varies between different samples as a result of their differing bulk composition, resulting in matrix-specific inaccuracy. We show how this can be accounted for via downhole fractionation corrections, resulting in an accuracy of better than 1% and a long-term reproducibility (intermediate precision) of <6% (relative standard deviation) in JCp-1NP using LA-ICP-MS in MR mode.
Conclusion: Our [K] measurement protocol is demonstrably precise and accurate and applicable to a wide range of materials. The measurement of K/Ca in relatively low-[K] marine carbonates is presented here as a key example of a new application opened up by these advances.

Der Roman lässt sich nicht nur als mit den Mitteln der Fiktion arbeitender Beitrag zur Geschichte der Verfolgung Schwarzer Menschen im Nationalsozialismus verstehen, sondern auch als selbst transatlantisches, vermeintlich historisches Zeitzeugnis, das aus einer ungewöhnlichen Perspektive die Thematik eines unsteten Archivs verflochtener Gewalt- und Exilerfahrungen beleuchtet. Das Augenmerk der Analyse liegt einerseits auf den Chancen eines solchen Archivs: Der Roman zeigt Möglichkeiten diasporischer Gemeinschaftsbildung und beschreibt die transatlantische Perspektive des Protagonisten als Ressource für das Erkennen von Kontinuitäten der Unterdrückung wie auch für das Ableiten von alltäglichen Widerstandsstrategien. Andererseits werden ausgehend vom Kunstgriff der Herausgeberfiktion und von Dokumenten aus dem Nachlass Risiken und Widersprüche des hier entworfenen Archivs diskutiert. Es wird gezeigt, dass dieses zwar auf Transnationalität zielt, jedoch teilweise der Rückbindung an ein lokales Moment bedarf.
Non-matrix-matched calibration of laser ablation ICPMS (trace/major) element data is a common quantification strategy. However, LA sampling is associated with downhole elemental fractionation, potentially causing inaccuracies if the magnitude of fractionation between the sample and reference material (RM) differs. Here, we estimate fractionation factors (FFs) for different elements (El) in a range of RMs relative to NIST SRM610/612 (FFEl/Ca-NIST) and evaluate element-specific corrections for downhole fractionation using these measured FFEl/Ca-NIST. Significantly different mean El/Ca values were observed before and after correction, particularly for the alkali elements (all RMs), and B, Fe, and Zn (some RMs), notably improving accuracy, especially for the alkali elements. In cases where this methodology does not result in an accuracy improvement, this may help identify underlying issues in reported/reference values for RMs, given that this phenomenon should be accounted for. Overall, we recommend considering routine assessment of FFs and applying a FF correction to enhance data quality.
Der Natrium-abhängige Kaliumkanal Slack (KNa1.1, Slo2.2, KCNT1) nimmt eine Schlüsselrolle in der Regulation neuronaler Erregbarkeit ein, indem er die Ausbildung und Feuerungsfrequenz von Aktionspotentialen kontrolliert. Sowohl in Mäusen als auch in Menschen wird Slack besonders hoch in nicht-peptidergen C-Faser-Neuronen exprimiert. Wissenschaftliche Erkenntnisse der letzten Jahre konnten die Beteiligung von Slack-Kanälen in der Signalverarbeitung neuropathischer Schmerzen, aber auch in verschiedenen Arten von Pruritus, feststellen. Dabei zeigen Slack-defiziente Mäuse ein verstärktes mechanisches Schmerzverhalten nach einer peripheren Nervenverletzung und ein erhöhtes Kratzverhalten in akuten Juckreiz-Modellen. Das als Slack-Aktivator identifizierte trizyklische Neuroleptikum Loxapin zeigt sowohl analgetische als auch antipruritische Effekte in Mäusen, jedoch ist sein klinischer Einsatz auf Grund schwerwiegender antipsychotischer Nebenwirkungen limitiert. Basierend auf Loxapins Leitstruktur wurden daher in dieser Arbeit neue Slack-Aktivatoren mit einem verbesserten pharmakologischen Profil designed und ihr Potential für die Therapie von Schmerzen sowie akutem und chronischem Pruritus in vivo untersucht.
The hepatitis B virus is one of the most common causes of virus-related chronic liver disease and remains a major global health problem affecting 296 million people worldwide. Despite an available and highly effective vaccination, hepatitis B infections lead to an annual mortality rate of approximately 0.8 million people. The global prevalence is heterogeneously distributed and reflects a high infections and chronicity, particularly in low-income countries, due to a lack of vaccination strategies, underdiagnosis and low treatment rates. A complete cure remains undiscovered to this day. Based on their genetic makeup, the virus is categorized into nine genotypes with a genetic difference of more than 8% within the sequence. In addition to their geographical distribution, hepatitis B virus genotypes also differ in terms of their clinical outcome, pathogenesis and treatment response.
The viral protein HBx is known to interact with several cellular signaling pathways and is thereby accounted as the driving force in the development of hepatitis B virus-associated pathogenesis and progression of hepatocellular carcinoma. In particular, HBx interacts with mitochondria and induces profound alterations in the mitochondrial morphology and function with a severe impact on the liver’s physiology and with an emerging role in liver-related disease progression.
This study aims to investigate the genotype-related impact of HBx with regard to their interaction with cellular signaling pathways. A particular focus was placed on mitochondria-dependent interactions and signaling pathways in order to broaden the understanding of the genetic diversity of the genotypes.
Differences between genotypes of HBx were examined and compared through in vitro experiments based on a cell culture-based system. Plasmid DNA encoding the HBx protein of the different genotypes was transiently transfected into Huh7 or HepG2 cells and examined for molecular and protein-biochemical effects on the host cell, usually 72 hours after transfection. This study focused on the most common genotypes A, B, C, D, E and G worldwide.
Based on initial kinome profiling analyses, it was found that HBx differs greatly within their genetic variants and suggests different effects on overall cell function and in particular on mitochondrial kinases. Furthermore, confocal laser scanning microscopy reveals profound HBx-mediated changes in the mitochondrial network structure, however with major differences among the different genotypes. In particular, HBx of genotypes A and G causes enormous fragmentation of mitochondrial structures, accompanied by emergent changes in mitochondrial function. Due to an increased interaction with the voltage-dependent anion channel 3, a significant loss of mitochondrial membrane potential was also observed, together with an increased radial oxygen stress level and an induction of central mitochondria-dependent inflammatory mediators. In contrast, the contribution of HBx-genotype B and E reveals only moderate effects in these regards. Using a pH-sensitive reporter system, HBx genotypes which previously indicated a strong distribution in the mitochondrial morphology and function, also showed an elevated mitophagy through the PINK1/Parkin-mediated pathway. This study provides direct evidence that HBx-mediated changes in host cell signaling pathways, especially in mitochondrial-associated pathways, fundamentally dependent on the different genotypes. In addition, the results also indicate an important role of HBx in the process of genotype-dependent liver pathogenesis and provide insight into the underlying cellular mechanisms and signaling pathways.

In vier Studien werden jüdische Erfahrungen der Moderne zwischen Zeigen und Beobachten, sozialer Schau und antisemitischer Bedrängnis als Modi des Theatralen zum Thema. Dabei rückt Theater als Praxis ins Zentrum, mit der Zugehörigkeit in der Moderne verhandelt, selbstbewusst behauptet oder befragt wurde. Während sich Juden und Jüdinnen "wie auf einer Bühne" permanenter Beobachtung ausgesetzt sahen, kommentierten sie dies wiederum theatral, erprobten eigene Strategien sozialer Teilhabe und beteiligten sich an der Ausgestaltung von bürgerlichen Sozialrollen in der Großstadt. Theater zeigt sich so als vielschichtige kulturelle Praxis wie als "Spiel- und Spiegelform des Lebens" (Stefan Zweig).
Stimulated emission depletion (STED) microscopy is a super-resolution technique that surpasses the diffraction limit and has contributed to the study of dynamic processes in living cells. However, high laser intensities induce fluorophore photobleaching and sample phototoxicity, limiting the number of fluorescence images obtainable from a living cell. Here, we address these challenges by using ultra-low irradiation intensities and a neural network for image restoration, enabling extensive imaging of single living cells. The endoplasmic reticulum (ER) was chosen as the target structure due to its dynamic nature over short and long timescales. The reduced irradiation intensity combined with denoising permitted continuous ER dynamics observation in living cells for up to 7 hours with a temporal resolution of seconds. This allowed for quantitative analysis of ER structural features over short (seconds) and long (hours) timescales within the same cell, and enabled fast 3D live-cell STED microscopy. Overall, the combination of ultra-low irradiation with image restoration enables comprehensive analysis of organelle dynamics over extended periods in living cells.
Correlative dynamic imaging of cellular landmarks, such as nuclei and nucleoli, cell membranes, nuclear envelope and lipid droplets is critical for systems cell biology and drug discovery, but challenging to achieve with molecular labels. Virtual staining of label-free images with deep neural networks is an emerging solution for correlative dynamic imaging. Multiplexed imaging of cellular landmarks from scattered light and subsequent demultiplexing with virtual staining leaves the light spectrum for imaging additional molecular reporters, photomanipulation, or other tasks. Current approaches for virtual staining of landmark organelles are fragile in the presence of nuisance variations in imaging, culture conditions, and cell types. We report training protocols for virtual staining of nuclei and membranes robust to variations in imaging parameters, cell states, and cell types. We describe a flexible and scalable convolutional architecture, UNeXt2, for supervised training and self-supervised pre-training. The strategies we report here enable robust virtual staining of nuclei and cell membranes in multiple cell types, including human cell lines, neuromasts of zebrafish and stem cell (iPSC)-derived neurons, across a range of imaging conditions. We assess the models by comparing the intensity, segmentations, and application-specific measurements obtained from virtually stained and experimentally stained nuclei and cell membranes. The models rescue missing labels, non-uniform expression of labels, and photobleaching. We share three pre-trained models (VSCyto3D, VSNeuromast, and VSCyto2D) and a PyTorch-based pipeline (VisCy) for training, inference, and deployment that leverages current community standards for image data and metadata.

In the ancient microbial Wood-Ljungdahl pathway, CO2 is fixed in a multi-step process with acetyl-CoA synthesis at the bifunctional carbon monoxide dehydrogenase/acetyl-CoA synthase complex (CODH/ACS). Here, we present catalytic snapshots of the CODH/ACS from the gas-converting acetogen Clostridium autoethanogenum, characterizing the molecular choreography of the overall reaction including electron transfer to the CODH for CO2 reduction, methyl transfer from the corrinoid iron-sulfur protein (CoFeSP) partner to the ACS active site and acetyl-CoA production. Unlike CODH, the multidomain ACS undergoes large conformational changes to form an internal connection to the CODH active site, accommodate the CoFeSP for methyl transfer and protect the reaction intermediates. Altogether, the structures allow us to draw a detailed reaction mechanism of this enzyme crucial for CO2 fixation in anaerobic organisms.
By analyzing (27.12±0.14)×108 ψ(3686) events accumulated with the BESIII detector, the decay ηc(2S)→K+K−η is observed for the first time with a significance of 6.2σ after considering systematic uncertainties. The product of the branching fractions of ψ(3686)→γηc(2S) and ηc(2S)→K+K−η is measured to be B(ψ(3686)→γηc(2S))×B(ηc(2S)→K+K−η)=(2.39±0.32±0.34)×10−6, where the first uncertainty is statistical, and the second one is systematic. The branching fraction of ηc(2S)→K+K−η is determined to be B(ηc(2S)→K+K−η)=(3.42±0.46±0.48±2.44)×10−3, where the third uncertainty is due to the branching fraction of ψ(3686)→γηc(2S). Using a recent BESIII measurement of B(ηc(2S)→K+K−π0), we also determine the ratio between the branching fractions of ηc(2S)→K+K−η and ηc(2S)→K+K−π0 to be 1.49±0.22±0.25, which is consistent with the previous result of BaBar at a comparable precision level.
Search for X(3872)→π⁰π⁰χc₁,₂

Using 10.1 fb−1 of e+e− collision data collected by the BESIII detector with center-of-mass energies between 4.15 GeV and 4.30 GeV, we search for the decays X(3872)→π0π0χc1,2, where the X(3872) is produced in e+e−→γX(3872). No evidence above 3σ is found for either decay. Upper limits at the 90% C.L. on the branching fractions of X(3872)→π0π0χc1,2 normalized to the branching fraction of X(3872)→π+π−J/ψ are set to be B(X(3872)→π0π0χc1)/B(X(3872)→π+π−J/ψ)<1.1 and B(X(3872)→π0π0χc2)/B(X(3872)→π+π−J/ψ)<0.5, taking into account both statistical and systematic uncertainties.

This work opens the possibility to investigate the conditions under which topological states develop in this series of strongly correlated 3-4-3 materials.

Heterostructures of graphene in proximity to magnetic insulators open the possibility to investigate exotic states emerging from the interplay of magnetism, strain and charge transfer between the layers. Recent reports on the growth of self-integrated atomic wires of β-RuCl3 on graphite suggest these materials as versatile candidates to investigate these effects. Here we present detailed first principles calculations on the charge transfer and electronic structure of β-RuCl3/heterostructures and provide a comparison with the work function analysis of the related honeycomb family members α-RuX3 (X = Cl,Br,I). We find that proximity of the two layers leads to a hole-doped graphene and electron-doped RuX3 in all cases, which is sensitively dependent on the distance between the two layers. Furthermore, strain effects due to lattice mismatch control the magnetization which itself has a strong effect on the charge transfer. Charge accumulation in β-RuCl3 strongly drops away from the chain making such heterostructures suitable candidates for sharp interfacial junctions in graphene-based devices.
Antimatter is a research topic of fundamental interest. Sufficient matter-antimatter asymmetry in the early Universe created the matter-dominated world today. The origin of this asymmetry is not completely understood to date. High-energy nuclear collisions create conditions similar to the Universe microseconds after the Big Bang, with comparable amounts of matter and antimatter. Much of the antimatter created escapes the rapidly expanding fireball without annihilation, making such collisions an effective experimental tool to create heavy antimatter nuclear objects and study their properties. In this paper, we report the first observation of the antimatter hypernucleus 4Λ¯H¯¯¯¯, composed of an Λ¯, an antiproton and two antineutrons. The discovery was made through its two-body decay after production in ultrarelativistic heavy-ion collisions by the STAR experiment at the Relativistic Heavy Ion Collider. In total, 15.6 candidate 4Λ¯H¯¯¯¯ antimatter hypernuclei are obtained with an estimated background count of 6.4. Lifetimes of the antihypernuclei 3Λ¯H¯¯¯¯ and 4Λ¯H¯¯¯¯ are measured and compared with the lifetimes of their corresponding hypernuclei, testing the symmetry between matter and antimatter. Various production yield ratios among (anti)hypernuclei and (anti)nuclei are also measured and compared with theoretical model predictions, shedding light on their production mechanism.

canning tunneling microscopy (STM) is perhaps the most promising way to detect the superconducting gap size and structure in the canonical unconventional superconductor Sr2RuO4 directly. However, in many cases, researchers have reported being unable to detect the gap at all in simple STM conductance measurements. Recently, an investigation of this issue on various local topographic structures on a Sr-terminated surface found that superconducting spectra appeared only in the region of small nanoscale canyons, corresponding to the removal of one RuO surface layer. Here, we analyze the electronic structure of various possible surface structures using first principles methods, and argue that bulk conditions favorable for superconductivity can be achieved when removal of the RuO layer suppresses the RuO4 octahedral rotation locally. We further propose alternative terminations to the most frequently reported Sr termination where superconductivity surfaces should be observed.
Motivated by recently reported magnetic-field induced topological phases in ultracold atoms and correlated Moiré materials, we investigate topological phase transitions in a minimal model consisting of interacting spinless fermions described by the Hofstadter model on a square lattice. For interacting lattice Hamiltonians in the presence of a commensurate magnetic flux it has been demonstrated that the quantized Hall conductivity is constrained by a Lieb-Schultz-Mattis (LSM)-type theorem due to magnetic translation symmetry. In this work, we revisit the validity of the theorem for such models and establish that a topological phase transition from a topological to a trivial insulating phase can be realized but must be accompanied by spontaneous magnetic translation symmetry breaking caused by charge ordering of the spinless fermions. To support our findings, the topological phase diagram for varying interaction strength is mapped out numerically with exact diagonalization for different flux quantum ratios and band fillings using symmetry indicators. We discuss our results in the context of the LSM-type theorem.
On the potential for GWAS with phenotypic population means and allele-frequency data (popGWAS)

This study explores the potential of a novel genome-wide association study (GWAS) approach for identifying loci underlying quantitative polygenic traits in natural populations. Extensive population genetic forward simulations demonstrate that the approach is generally effective for oligogenic and moderately polygenic traits and relatively insensitive to low heritability, but applicability is limited for highly polygenic architectures and pronounced population structure. The required sample size is moderate with very good results being obtained already for a few dozen populations scored. The method performs well in predicting population means even with a moderate false positive rate. When combined with machine learning for feature selection, this rate can be further reduced. The data efficiency of the method, particularly when using pooled sequencing, makes GWAS studies more accessible for research in biodiversity genomics. Overall, this study highlights the promise of this popGWAS approach for dissecting the genetic basis of complex traits in natural populations.
Abstract
Seed harvesting from wild plant populations is key for ecological restoration, but may threaten the persistence of source populations. Consequently, several countries have set guidelines limiting the proportions of harvestable seeds. However, these guidelines are so far inconsistent, and they lack a solid empirical basis. Here, we use high-resolution data from 298 plant species to model the demographic consequences of seed harvesting. We find that the current guidelines do not protect populations of annuals and short-lived perennials, while they are overly restrictive for long-lived plants. We show that the maximum possible fraction of seed production – what can be harvested without compromising the long-term persistence of populations – is strongly related to the generation time of the target species. When harvesting every year, this safe seed fraction ranges from 80% in long-lived species to 2% in most annuals. Less frequent seed harvesting substantially increases the safe seed fraction: In the most vulnerable annual species, it is safe to harvest 5%, 10% or 30% of population seed production when harvesting every two, five or ten years, respectively. Our results provide a quantitative basis for seed harvesting legislations worldwide, based on species’ generation time and harvesting regime.
Significance The UN Decade on Ecosystem Restoration, 2021-2030, foresees upscaling restoration, and the demand for native seed is skyrocketing. Seeds for restoring native vegetation are often harvested in wild, but too intensive harvest can threaten the donor populations. Existing guidelines that set limits to wild seed harvest are mostly based on expert opinions, yet they commonly lack empirical basis and vary among regions in one order of magnitude. We show that the current guidelines urgently need to be reformulated, because they are overly restrictive in long-lived species, while they do not protect annual plants from extinction. Using matrix population models of nearly 300 plant species, we provide a quantitative basis for a new seed harvesting legislation world-wide.
Evidence for the singly Cabibbo suppressed decay Λ+c→pπ0 is reported for the first time with a statistical significance of 3.7σ based on 6.0 fb−1 of e+e− collision data collected at center-of-mass energies between 4.600 and 4.843 GeV with the BESIII detector at the BEPCII collider. The absolute branching fraction of Λ+c→pπ0 is measured to be (1.56+0.72−0.58±0.20)×10−4. Combining with the branching fraction of Λ+c→nπ+, (6.6±1.3)×10−4, the ratio of the branching fractions of Λ+c→nπ+ and Λ+c→pπ0 is calculated to be 3.2+2.2−1.2. As an important input for the theoretical models describing the decay mechanisms of charmed baryons, our result indicates that the non-factorizable contributions play an essential role and their interference with the factorizable contributions should not be significant. In addition, the absolute branching fraction of Λ+c→pη is measured to be (1.63±0.31stat±0.11syst)×10−3.
We measure triangular flow relative to the reaction plane at 3 GeV center-of-mass energy in Au+Au collisions at the BNL Relativistic Heavy Ion Collider. A significant v3 signal for protons is observed, which increases for higher rapidity, higher transverse momentum, and more peripheral collisions. The triangular flow is essentially rapidity-odd with a slope at mid-rapidity, dv3/dy|(y=0), opposite in sign compared to the slope for directed flow. No significant v3 signal is observed for charged pions and kaons. Comparisons with models suggest that a mean field potential is required to describe these results, and that the triangular shape of the participant nucleons is the result of stopping and nuclear geometry.
The Born cross sections for the process e+e−→ωη′ are measured at 22 center-of-mass energies from 2.000 to 3.080 GeV using data collected with the BESIII detector at the BEPCII collider. A resonant structure is observed with a statistical significance of 9.6σ. A Breit-Wigner fit determines its mass to be MR=(2153±30±31) MeV/c2 and its width to be ΓR=(167±77±7) MeV, where the first uncertainties are statistical and the second are systematic.
Using a sample of (10087±44)×106 J/ψ events, which is about 45 times larger than that was previously analyzed, a further investigation on the J/ψ→γ3(π+π−) decay is performed. A significant distortion at 1.84 GeV/c2 in the line-shape of the 3(π+π−) invariant mass spectrum is observed for the first time, which could be resolved by two overlapping resonant structures, X(1840) and X(1880). The new state X(1880) is observed with a statistical significance larger than 10σ. The mass and width of X(1880) are determined to be 1882.1±1.7±0.7 MeV/c2 and 30.7±5.5±2.4 MeV, respectively, which indicates the existence of a pp¯ bound state.
Using 9.0 fb−1 of e+e− collision data collected at center-of-mass energies from 4.178 to 4.278 GeV with the BESIII detector at the BEPCII collider, we perform the first search for the radiative transition χc1(3872)→γψ2(3823). No χc1(3872)→γψ2(3823) signal is observed. The upper limit on the ratio of branching fractions B(χc1(3872)→γψ2(3823),ψ2(3823)→γχc1)/B(χc1(3872)→π+π−J/ψ) is set as 0.075 at the 90\% confidence level. Our result contradicts theoretical predictions under the assumption that the χc1(3872) is the pure charmonium state χc1(2P).
Using data samples with an integrated luminosity of 22.42 fb−1 collected by the BESIII detector operating at the BEPCII storage ring, we measure the cross sections of the 𝑒+𝑒−→𝜂𝐽/𝜓 process at center-of-mass energies from 3.808 to 4.951 GeV. Three structures are observed in the line shape of the measured cross sections. A maximum-likelihood fit with 𝜓(4040), two additional resonances, and a nonresonant component are performed. The mass and width of the first additional state are (4219.7±2.5±4.5) MeV/𝑐2 and (80.7±4.4±1.4) MeV, respectively, consistent with the 𝜓(4230). For the second state, the mass and width are (4386±13±17) MeV/𝑐2 and (177±32±13) MeV, respectively, consistent with the 𝜓(4360). The first uncertainties are statistical, and the second ones are systematic. The statistical significance of 𝜓(4040) is 8.0𝜎 and those for 𝜓(4230) and 𝜓(4360) are more than 10.0𝜎.
We present the first observation of the singly Cabibbo-suppressed decay Λ+c→ΛK+π0 with a significance of 5.7σ and the first evidence of Λ+c→ΛK+π+π− decay with a significance of 3.1σ, based on e+e− annihilation data recorded by the BESIII detector at the BEPCII collider. The data correspond to an integrated luminosity of 6.4 fb−1, in the center-of-mass energy range from 4.600 GeV to 4.950 GeV. We determine the branching fractions of Λ+c→ΛK+π0 and Λ+c→ΛK+π+π− relative to their Cabibbo-favored counterparts to be B(Λ+c→ΛK+π0)B(Λ+c→Λπ+π0)=(2.09±0.39stat.±0.07syst.)×10−2 and B(Λ+c→ΛK+π+π−)B(Λ+c→Λπ+π+π−)=(1.13±0.41stat.±0.06syst.)×10−2, respectively. Moreover, by combining our measured result with the world average of B(Λ+c→Λπ+π0), we obtain the branching fraction B(Λ+c→ΛK+π0)=(1.49±0.27stat.±0.05syst.±0.08ref.)×10−3. This result significantly departs from theoretical predictions based on quark SU(3) flavor symmetry, which is underpinned by the presumption of meson pair S-wave amplitude dominance.
Using e+e− annihilation data corresponding to an integrated luminosity of 2.93 fb−1 taken at the center-of-mass energy s√=3.773~GeV with the BESIII detector, a joint amplitude analysis is performed on the decays D0→π+π−π+π− and D0→π+π−π0π0(non-η). The fit fractions of individual components are obtained, and large interferences among the dominant components of D0→a1(1260)π, D0→π(1300)π, D0→ρ(770)ρ(770) and D0→2(ππ)S are found in both channels. With the obtained amplitude model, the CP-even fractions of D0→π+π−π+π− and D0→π+π−π0π0(non-η) are determined to be (75.2±1.1stat.±1.5syst.)% and (68.9±1.5stat.±2.4syst.)%, respectively. The branching fractions of D0→π+π−π+π− and D0→π+π−π0π0(non-η) are measured to be (0.688±0.010stat.±0.010syst.)% and (0.951±0.025stat.±0.021syst.)%, respectively. The amplitude analysis provides an important model for binning strategy in the measurements of the strong phase parameters of D0→4π when used to determine the CKM angle γ(ϕ3) via the B−→DK− decay.

Observation of η_(c)(1S, 2S) and χ_(cJ) decays to 2(π⁺π^(−))η via ψ(3686) radiative transitions

Based on 2.7×109 ψ(3686) decays collected with the BESIII detector, the radiative decay ψ(3686)→γ2(π+π−)η is investigated to measure properties of S- and P-wave charmonium states. The branching fraction of the decay ηc(1S)→2(π+π−)η, which is found to have a strong dependence on the interference pattern between ηc(1S) and non-ηc(1S) processes, is measured in both destructive and constructive interference scenarios for the first time. The mass and width of the ηc(1S) are measured to be M=(2984.14±0.13±0.38) MeV/c2 and Γ=(28.82±0.11±0.82) MeV, respectively. Clear signals for the decays of the χcJ(J=0,1,2) and the ηc(2S) to 2(π+π−)η are also observed for the first time, and the corresponding branching fractions are measured. The ratio of the branching fractions between the ηc(2S) and ηc(1S) decays is significantly lower than the theoretical prediction, which might suggest different dynamics in their decays.
A light scalar X0 or vector X1 particles have been introduced as a possible explanation for the (g−2)μ anomaly and dark matter phenomena.
Using (8.998±0.039)×109 $\jpsi$ events collected by the BESIII detector, we search for a light muon philic scalar X0 or vector X1 in the processes J/ψ→μ+μ−X0,1 with X0,1 invisible decays. No obvious signal is found, and the upper limits on the coupling g′0,1 between the muon and the X0,1 particles are set to be between 1.1×10−3 and 1.0×10−2 for the X0,1 mass in the range of 1<M(X0,1)<1000 MeV/c2 at 90% confidence level.
Using 2.93 fb−1 of e+e− collision data collected with the BESIII detector at the center-of-mass energy of 3.773 GeV, we investigate the semileptonic decays D+→π+π−ℓ+νℓ (ℓ=e and μ). The D+→f0(500)μ+νμ decay is observed for the first time. By analyzing simultaneously the differential decay rates of D+→f0(500)μ+νμ and D+→f0(500)e+νe in different ℓ+νℓ four-momentum transfer intervals, the product of the relevant hadronic form factor ff0+(0) and the magnitude of the c→d Cabibbo-Kobayashi-Maskawa matrix element |Vcd| is determined to be ff0+(0)|Vcd|=0.0787±0.0060stat±0.0033syst for the first time. With the input of |Vcd| from the global fit in the standard model, we determine ff0+(0)=0.350±0.027stat±0.015syst. The absolute branching fractions of D+→f0(500)(π+π−)μ+νμ and D+→ρ0(π+π−)μ+νμ are determined as (0.72±0.13stat±0.10syst)×10−3 and (1.64±0.13stat±0.11syst)×10−3. Combining these results with those of previous BESIII measurements on their semielectronic counterparts from the same data sample, we test lepton flavor universality by measuring the branching fraction ratios BD+→ρ0μ+νμ/BD+→ρ0e+νe = 0.88±0.10 and BD+→f0(500)μ+νμ/BD+→f0(500)e+νe = 1.14±0.28, which are compatible with the standard model expectation.
Using 7.33~fb−1 of e+e− collision data collected by the BESIII detector at center-of-mass energies in the range of s√=4.128−4.226~GeV, we search for the rare decays D+s→h+(h0)e+e−, where h represents a kaon or pion. By requiring the e+e− invariant mass to be consistent with a ϕ(1020), 0.98<M(e+e−)<1.04 ~GeV/c2, the decay D+s→π+ϕ,ϕ→e+e− is observed with a statistical significance of 7.8σ, and evidence for the decay D+s→ρ+ϕ,ϕ→e+e− is found for the first time with a statistical significance of 4.4σ. The decay branching fractions are measured to be B(D+s→π+ϕ,ϕ→e+e−)=(1.17+0.23−0.21±0.03)×10−5, and B(D+s→ρ+ϕ,ϕ→e+e−)=(2.44+0.67−0.62±0.16)×10−5, where the first uncertainties are statistical and the second systematic. No significant signal for the three four-body decays of D+s→π+π0e+e−, D+s→K+π0e+e−, and D+s→K0Sπ+e+e− is observed. For D+s→π+π0e+e−, the ϕ mass region is vetoed to minimize the long-distance effects. The 90% confidence level upper limits set on the branching fractions of these decays are in the range of (7.0−8.1)×10−5.
Based on (2.712±0.014)×109 ψ(3686) events collected by the BESIII collaboration, evidence of the hadronic decay hc→K0SK+π−+c.c. is found with a significance of 4.3σ in the ψ(3686)→π0hc process. The branching fraction of hc→K0SK+π−+c.c. is measured to be (7.3±0.8±1.8)×10−4, where the first and second uncertainties are statistical and systematic, respectively. Combining with the exclusive decay width of ηc→KK¯π, our result indicates inconsistencies with both pQCD and NRQCD predictions.
The epitranscriptome embodies many new and largely unexplored functions of RNA. A major roadblock in the epitranscriptomics field is the lack of transcriptome-wide methods to detect more than a single RNA modification type at a time, identify RNA modifications in individual molecules, and estimate modification stoichiometry accurately. We address these issues with CHEUI (CH3 (methylation) Estimation Using Ionic current), a new method that concurrently detects N6-methyladenosine (m6A) and 5-methylcytidine (m5C) in individual RNA molecules from the same sample, as well as differential methylation between any two conditions. CHEUI processes observed and expected nanopore direct RNA sequencing signals with convolutional neural networks to achieve high single-molecule accuracy and outperforms other methods in detecting m6A and m5C sites and quantifying their stoichiometry. CHEUI’s unique capability to identify two modification types in the same sample reveals a non-random co-occurrence of m6A and m5C in mRNA transcripts in cell lines and tissues. CHEUI unlocks an unprecedented potential to study RNA modification configurations and discover new epitranscriptome functions.
While high-quality climate reconstructions of some past warm periods in the Cenozoic era now exist, the geological processes responsible for driving the observed longterm changes in atmospheric CO2 are not sufficiently well understood. The long-term change in atmospheric CO2 across the Cenozoic has been proposed to be driven by processes such as terrestrial weathering, organic carbon production and burial, reverse weathering, and volcanic degassing. One way of constraining the relative importance of the various driving forces proposed so far is to better understand the degree to which ocean chemistry has changed because the chemistry of seawater responds to geologic processes that drive atmospheric CO2. In addition, knowledge of the concentration of the major elements in seawater is crucial for accurately applying proxies such as those based on the boron isotopic composition and Mg/Ca of marine carbonates (a proxy for palaeo pH/CO2 and palaeotemperature, respectively). Previously reported records of seawater composition are primarily derived from fluid inclusions in marine evaporites; however, the results are sparse due to the limited availability of such deposits. In this thesis, changes in the Eocene seawater chemistry were reconstructed using trace element (elements/Ca) and isotopic (δ26Mg) proxies in a Larger Benthic Foraminifera (LBFs), i.e., Nummulites sp., to constrain the driving processes of long-term changes in seawater chemistry.
To achieve the objective of this thesis, first, a measurement protocol was established using LA-ICPMS to measure the K/Ca ratio simultaneously with other element/calcium ratios, which is challenging due to the interference of ArH+ on K+. Utilising this newly established measurement protocol, laboratory-cultured Operculina ammonoides grown at different seawater calcium concentrations ([Ca2+]), repeated at different temperatures, as well as modern O. ammonoides collected from different regions exhibiting a range of seawater parameters, were investigated. A significant correlation was observed between K/Casw and K/CaLBF, allowing K/CaLBF to potentially be used as a proxy for seawater major ion reconstructions. In addition, modern O. ammonoides demonstrated no significant influence of most seawater parameters (temperature, salinity, pH, or [CO32-]) on K/CaLBF. Modern O.
ammonoides were also assessed for their Mg isotopic composition (δ26Mg), revealing no significant effect of temperature or salinity on δ26MgLBF. Furthermore, the Mg isotopic fractionation in O. ammonoides was found to be close to that of inorganic calcite, indicating minimal vital effects in these large benthic foraminifera.
Operculina ammonoides is the nearest living relative of the abundant Eocene genus Nummulites, enabling the reconstruction of seawater chemistry using the calibration based on O. ammonoides. The trace elemental/calcium proxies, including Na/Ca, K/Ca, and Mg/Ca, as well as the δ26Mg proxy, were investigated in Eocene Nummulites. The result showed that during the Eocene, [Ca2+]sw was 1.6-2 times higher, while [K+]sw was ~2 times lower than the modern seawater composition. Furthermore, [Mg2+]sw decreased from the early Eocene (54.3− +9 7..69 mmol kg-1 at ~55 Ma) to Late Eocene (37.8− +4 4..3 4 mmol kg-1 at ~31 Ma), followed by
an increase toward modern seawater [Mg]. In contrast, the variability in δ26Mgsw values remained within a narrow range of ~0.3 ‰ throughout the Cenozoic. The reconstructed [Ca2+]sw agrees with the suggestion that Cenozoic seawater chemistry changes can be explained via a change in the seafloor spreading rate. When combined with existing records, the observed minimal change in δ26Mgsw with an increase in [Mg2+]sw suggests an additional possible role of a decrease in the formation of authigenic clay minerals coincident with the Cenozoic decline in deep ocean temperature, which is also supported by the increase in the [K+]sw reconstructed here for the first time. This finding highlights that the reduction in seafloor-spreading rate and decline in reverse weathering during the Cenozoic era has played a significant role in the evolution of seawater chemistry, emphasizing the importance of these processes in driving long-term changes in the carbon cycle.
Die Epoche der Weimarer Republik war gekennzeichnet durch eine Vielzahl von Umbrüchen im politischen, ökonomischen und gesellschaftlichen Bereich. Diese durch den Ersten Weltkrieg verursachten radikalen Veränderungen der Lebensverhältnisse hatten auch einen weitreichenden Einfluss auf den Alltag und die öffentliche Wahrnehmung der Frauen dieser Zeit: So konnten diese ab 1918 an den Wahlen teilnehmen, konnten sich an allen deutschen Universitäten einschreiben und in bestimmtem Maße finanziell unabhängig werden. Das Bild der so genannten "Neuen Frau", das gleichzeitig in der Öffentlichkeit generiert wurde, war insbesondere gekennzeichnet durch innerliche und äußerliche Emanzipation und stellte damit einen radikalen Gegenpol zur traditionellen Weiblichkeit des 19. Jahrhunderts dar. Viele Facetten der "Neuen Frau" sind in der Forschung weitgehend erschlossen - die sportliche Frau, die automobile Frau, die Frau als flapper oder garçonne - nur einer Ausprägung des neuen Frauentypus wurde bisher wenig Beachtung geschenkt: der Frau als Verbrecherin. Ziel der vorliegenden interdisziplinär und medienkomparatistisch angelegten Dissertation ist es daher, übergreifende Semantiken und Ästhetiken der geschlechtsspezifischen Verbrechensdarstellung in der Weimarer Republik offenzulegen und die unterschiedlich erzeugten, vermittelten und klassifizierenden Vorstellungen von Verbrecherinnen zu analysieren und zu kontextualisieren. Das zugrundeliegende Material umfasst dabei fachwissenschaftliche, publizistische und literarische Darstellungen von Verbrecherinnen, wobei das Hauptaugenmerk auf der Analyse von Artikeln aus Illustrierten Magazinen liegt, da diese, neben Radio und Film, zu den wirkungsmächtigsten Medien der Weimarer Republik gehören.
The PhD addresses the feasibility of reconstructing open charm mesons with the Compressed Baryonic Matter experiment, which will be installed at the FAIR accelerator complex at Darmstadt/Germany. The measurements will be carried out by means of a dedicated Micro Vertex Detector (MVD), which will be equipped with CMOS Monolithic Active Pixel Sensors (MAPS). The feasibility of reconstructing the particles with a proposed detector setup was studied.
To obtain conclusive results, the properties of a MAPS prototype were measured in a beam test at the CERN-SPS accelerator. Based on the results achieved, a dedicated simulation software for the sensors was developed and implemented into the software framework of CBM (CBMRoot). Simulations on the reconstruction of D0-mesons were carried out. It is concluded that the reconstruction of those particles is possible.
The PhD introduces the physics motivation of doing open charm measurements, represents the results of the measurements of MAPS and introduces the innovative simulation model for those sensors as much as the concept and results of simulations of the D0 reconstruction.
Why do banks issue contingent convertible debt? To answer this question we study comprehensive data covering all issues by publicly traded banks in Europe of contingent convertible bonds (CoCos) that count as additional tier 1 capital (AT1). We find that banks with lower asset volatility are more likely to issue AT1 CoCos than their riskier counterparts, but that CDS spreads do not react following issue announcements. Our estimates therefore suggest that agency costs play a crucial role in banks' ability to successfully issue CoCos. The agency costs may be higher for CoCos than for equity explaining why we observe riskier or lowly capitalized banks to issue equity rather than CoCos.
Over the last few decades, considerable attention has focused on small-scale studies of invasive plants and invaded systems. Unfortunately, small scale studies rarely provide comprehensive insight into the complexities of biological invasions at macroscales. Systematic and repeated monitoring of biological invasions at broad scales are rare. In this report, we highlight a unique invasive plant database from the national Forest Inventory and Analysis (FIA) program of the United States Forest Service. We demonstrate the importance and capability of this subcontinental-wide database by showcasing several critical macroscale invasion patterns that have emerged from its initial analysis: (1) large portion of the forests systems (39%) in the United States are impacted by invasive plants, (2) forests in the eastern United States harbor more invasive species than the western regions, (3) human land-use legacies at regional to national scalesb may drive large-scale invasion patterns. This accumulated dataset, which continues to grow in temporal richness with repeated measurements, will allow the understanding of invasion patterns and processes at multi-spatial and temporal scales. Such insights are not possible from smaller-scale studies, illustrating the benefit that can be gained by investing in the development of regional to continental-wide invasion monitoring programs elsewhere.
Ziel: Nebennierenraumforderungen können sonographisch mit guter Sensitivität und Spezifität erkannt werden. Ziel der vorliegenden Studie war es, die Wertig-keit der Kontrastmittel-Sonographie (CEUS = contrast enhanced ultrasound) unter Verwendung von Zeit-Intensitäts-Kurven für die Charakterisierung von Nebennierenraumforderungen zu bewerten.
Material und Methoden: 108 Patienten mit 116 Raumforderungen der Neben-niere erhielten einen Ultraschall der Nebenniere, inklusive einer Kontrastmittel-Sonographie mit dem Kontrastmittel SonoVue®. Die Kontrastmitteldynamik wurde mittels Zeit-Intensitätskurven aufgezeichnet. Anhand der Kontrastmittel-anflutungszeit wurden folgende vier Kontrastmittelanreicherungsmuster ver-wendet, welche in einer Pilotstudie1 entwickelt wurden: Anreicherungsmuster I = früh arterielle Kontrastmittelanreicherung, Anreicherungsmuster II = arterielle Kontrastmittelanreicherung, Anreicherungsmuster III = späte Kontrastmittelan-reicherung, Anreicherungsmuster IV = keine Kontrastmittelanreicherung. Zu-sätzlich erhielten alle Patienten eine Beurteilung der Nebenniere durch ein zweite Bildgebendes Verfahren (Computertomographie (CT) oder Magnetresonanztomographie (MRT)) sowie eine laborchemische Hormondiagnostik. In Fällen mit malignitätsverdächtigem Befund erfolgte eine Biopsie oder Adrenalektomie.
Ergebnisse: CEUS- Anreicherungsmuster I und II konnten bei allen Patienten mit primärem oder sekundärem Malignom der Nebenniere (n = 16) nachgewie-sen werden. Die Sensitivität der CEUS für die Diagnose einer malignen Neben-nierenraumforderung betrug 100% (95% CI [75; 100]) und die Spezifität 67% (95% CI [56; 75]). Bei 40 Nebennierenraumforderungen war die Histologie die Referenzmethode. In 68% dieser Nebennierenraumforderungen waren MRT/CT und CEUS kongruent in der Beurteilung der Dignität. Schlussfolgerung: Die Kontrastmittel-Sonographie ist eine nützliche ergän-zende Methode in der Aufarbeitung von Nebennierenraumforderungen mit exzellenter Sensitivität für die Diagnose eines Malignoms.
Most elements heavier than iron are synthesized in stars during neutron capture reactions in the r- and s-process. The s-process nucleosynthesis is composed of the main and weak component. While the s-process is considered to be well understood, further investigations using nucleosynthesis simulations rely on measured neutron capture cross sections as crucial input parameters. Neutron capture cross sections
relevant for the s-process can be measured using various experimental methods. A prominent example is the activation method relying on the 7Li(p,n)7Be reaction as a neutron source, which has the advantage of high neutron intensities and is able to create a quasi-stellar neutron spectrum at kBT = 25 keV. Other neutron sources able to provide quasi-stellar spectra at different energies suffer from lower neutron intensities. Simulations using the PINO tool suggest the neutron activation of samples with different neutron spectra, provided by the 7Li(p,n)7Be reaction, and a subsequent linear combination of the obtained spectrum-averaged cross sections
to determine the Maxwellian-averaged cross section (MACS) at various energies of astrophysical relevance. To investigate the accuracy of the PINO tool at proton energies between the neutron emission threshold at Ep = 1880.4 keV and 2800 keV,
measurements of the 7Li(p,n)7Be neutron fields are presented, which were carried out at the PTB Ion Accelerator Facility at the Physikalisch-Technische Bundesanstalt in Braunschweig. The neutron fields of ten different proton energies were measured.
The presented neutron fields show a good agreement at proton energies Ep = 1887, 1897, 1907, 1912 and 2100 keV. For the other proton energies, E p = 2000, 2200, 2300, 2500, and 2800 keV, differences between measurement and simulation were found and discussed. The obtained results can be used to benchmark and adapt the PINO tool and provide crucial information for further improvement of the neutron activation method for astrophysics.
An application for the 7Li(p,n)7Be neutron fields is presented as an activation experiment campaign of gallium, an element that is mostly produced during the weak s-process in massive stars. The available cross section data for the 69,71Ga(n,γ)
reactions, mostly determined by activation measurements, show differences up toa factor of three. To improve the data situation, activation measurements were carried out using the 7Li(p,n)7Be reaction. The neutron capture cross sections for
a quasi-stellar neutron spectrum at kBT = 25 keV were determined for 69Ga and 71Ga.
In this dissertation the formal abstraction and verification of analog circuit is examined. An approach is introduced that automatically abstracts a transistor level circuit with full Spice accuracy into a hybrid automaton (HA) in various output languages. The generated behavioral model exhibits a significant simulation speed-up compared to the original netlist, while maintaining an acceptable accuracy, and can be therefore used in various verification and validation routines. On top of that, the generated models can be formally verified against their Spice netlists, making the obtained models correct by construction.
The generated abstract models can be extended to enclose modeling as well as technology dependent parameter variations with little over approximations. As these models enclose the various behaviors of the sampled netlists, the obtained models are of significant importance as they can replace several simulations with just a single reachability analysis or symbolic simulation. Moreover, these models can be as well be used in different verification routines as demonstrated in this dissertation.
As the obtained models are described by HAs with linear behaviors in the locations, the abstract models can be as well compositionally linked, allowing thereby the abstraction of complex analog circuits.
Depending on the specified modeling settings, including for example the number of locations of the HA and the description of the system behavior, the accuracy, speedup, and various additional properties of the HA can be influenced. This is examined in detail in this dissertation. The underlying abstraction process is first covered in detail. Several extensions are then handled including the modeling of the HAs with parameter variations. The obtained models are then verified using various verification methodologies. The accuracy and speed-up of the abstraction methodology is finally evaluated on several transistor level circuits ranging from simple operational amplifiers up to a complex circuits.
Intelligenz ist einer der stärksten Einzelprädikatoren für berufliche Leistungen. Hochintelligente Personen, sogenannte Hochbegabte, müssten daher generell hohe berufliche Leistungen erbringen. Dies steht jedoch im Widerspruch zu Darstellungen von Schwierigkeiten Hochbegabter im Beruf. Sollten die negativen Stereotype zutreffen, wäre dies aus ressourcenorientierter Sicht problematisch, da Organisationen das der Hochbegabung zugrunde liegende Potenzial der Mitarbeiter*innen nicht hinreichend nutzen. Die vorliegende Arbeit leistet einen Beitrag dazu, diese Diskrepanz aufzuklären, indem sie untersucht wie Hochbegabte ihre berufliche Situation (erfolgreich) gestalten. Nach einer Begriffsklärung und Darstellung des Zusammenhangs von Intelligenz und Beruf wird im ersten Beitrag der internationale Forschungsstand mithilfe eines Literature Reviews systematisiert. Aufbauend darauf werden im zweiten Beitrag mithilfe einer Fragebogenerhebung die überwiegend internationalen Beiträge durch eine deutsche Stichprobe und die bisher wenig betrachtete Gruppe von Hochbegabten gemäß Kompetenzdefinition ergänzt. Schließlich fokussiert der dritte Beitrag den Widerspruch, dass Hochbegabte generell erfolgreich und zufrieden im Beruf sind, jedoch individuell von Schwierigkeiten berichten. Dazu werden teilstrukturierte Interviews durchgeführt, um ein tieferes Verständnis für den Einfluss der Hochbegabung auf den Beruf sowie etwaige bestehende Unterschiede zu nicht-hochbegabten Personen zu gewinnen. Zudem werden Strategien erfasst, die Hochbegabte nutzen, um Unterschiede und eventuell daraus resultierende Schwierigkeiten zu bewältigen. Abschließend werden Implikationen für die Berufspraxis und Forschung diskutiert.
Mast cells are long-lived tissue-resident leukocytes, located most abundantly in the skin and mucosal surfaces. They belong to the first line of defence of the body, protecting against invading pathogens, toxins and allergens. Their secretory granules are densely packed with a plethora of mediators, which can be released immediately upon activation of the cell. Next to their role in IgE-mediated allergic diseases and in promoting inflammation, potential anti-inflammatory functions have been assigned to mast cells, depending on the biological setting. The aim of this thesis was to contribute to a better understanding of the role of mast cells during the resolution of a local inflammation. Therefore, in a first of step a suitable model of a local inflammation had to be identified. Since comparison of the two Toll-like receptor (TLR)-agonists zymosan and lipopolysaccharide (LPS), which are most commonly used to locally induce inflammation, revealed a systemic response after LPS-injection and a local inflammation after zymosan-injection, the TLR2 agonist zymosan was chosen for the subsequent experiments. Multi epitope ligand cartography (MELC) combined with statistical neighbourhood analysis showed that mast cells are located in an anti-inflammatory microenvironment next to M2 macrophages during resolution of inflammation, while neutrophils and M1 macrophages are located in the zymosan-filled core of the inflammation. Furthermore, infiltrating neutrophils during peak inflammation and an increasing population of macrophages phagocytosing neutrophils during resolution of inflammation could be observed. MELC as well as flow cytometry analysis of mast cell-deficient mice revealed a decreased phagocytosing activity of macrophages in the absence of mast cells. As an untargeted approach to identify mast cell-derived mediators induced by zymosan, mRNA sequencing of bone marrow-derived mast cells (BMMCs) was performed. Gene ontology term analysis of the sequencing data revealed the induction of the type I interferon (IFN) pathway as the dominant response. Contradicting previous studies, I could validate the production of IFN-β by mast cells in response to zymosan and LPS in vitro. Furthermore IFN-β expression by mast cells was also detected in vivo. In accordance with previous studies regarding other cell types the release of IFN-β by mast cells depends on endosomal signaling. The potential of IFN-β to enhance the phagocytosing activity of macrophages has been demonstrated recently. Besides IFN-β, various other mediators with reported enhancing effects on macrophage phagocytosis were also induced by zymosan in BMMCs, including Interleukin (IL)-1β, IL-4, IL-13, and Prostaglandin (PG) E2. Thus, either one of these mediators alone or a combination of them could promote macrophage phagocytosis.
In conclusion, I herein present mast cells as a novel source for IFN-β induced by non-viral TLR ligands and demonstrate their enhancing effect on macrophage phagocytosis, thereby contributing to the resolution of inflammation.

Im Vordergrund der Betrachtungen steht die Inszenierung einer homosexuellen Beziehung zweier Frauen in den 1950er Jahren, die bereits in der literarischen Vorlage, die 1952 unter dem Titel "The Price of Salt" erschien, in den Termini der aristotelisch geprägten Tragödientradition gesprochen, nicht in einer Katastrophe mündet, sondern ein (stilles) Happy End für die Protagonistinnen vorsieht. Polek stellt heraus, dass der im Jahr 2015 erschienene Film nicht nur die Thematisierung von Homosexualität vornimmt, sondern diese vor allem mit den damit kollidierenden Lebensentwürfen verbindet, die in den gesellschaftlichen Verhältnissen im nordamerikanischen Raum der 1950er Jahre vorherrschten. Dies gelingt dem Regisseur Todd Haynes im Medium des Films durch formale Gestaltungsmittel in Form der Montage sowie der visuellen Darstellung geschlechtsspezifischer Räume, die eine entscheidende Bedeutung für die Veränderung und Dekonstruktion scheinbar stabiler Entwürfe der Geschlechterrollen einnehmen.
Die vorliegende Arbeit mit dem Titel Multiphoton Processes and Photocontrol of Biochemical Reaction Pathways befasst sich mit verschiedenen Strategien zur Implementierung von optischer Kontrolle in biochemisch relevanten Systemen. Auf systemischer Ebene wurde einerseits die Licht-getriebene Natriumpumpe Krokinobacter Eikastus Rhodopsin 2 (KR2) vor dem Hintergrund optogenetischer Anwendungen untersucht, und andererseits die Optimierung der wichtigsten photochemischen Parameter von photolabilen Schutzgruppen (PPG, engl. photolabile protecting group) angestrebt. Von der technisch-photophysikalischen Seite wurde ein weiterer Fokus auf den Aktivierungs- und Deaktivierungsschritt gelegt. Hierbei wurden vor allem Mehrphotonen-Prozesse betrachtet, die entweder durch simultane Absorption zweier Photonen zu einer spezifischen hoch-energetischen Anregung führen, oder durch sequentielle Absorption eine sukzessive Aktivierung und Deaktivierung eines Systems bewerkstelligen können. Auch wenn der Schwerpunkt dieser schriftlichen Arbeit auf den spektroskopischen Ergebnissen liegt, waren alle hier diskutierten Projekte stark kollaborativ und umfassten eine große Bandbreite verschiedener Techniken. Dies spiegelt den interdisziplinären Charakter vieler aktueller Fragestellungen in der photochemischen Forschung wider, die - in vielen Fällen - letztlich auf medizinische oder pharmazeutische Fortschritte abzielen.
Zunächst wurde die lichtgetriebene Natriumpumpe KR2 untersucht, die durch ihre mögliche Anwendung als optogenetisches Werkzeug bekannt wurde. In einer vergleichenden Studie der Natrium- und Protonenpumpmodi von KR2 konnten wichtige mechanistische Prinzipien für die Funktionalität des Proteins identifiziert werden. Dazu gehört die direkte Beteiligung spezifischer Strukturmerkmale wie die Aminosäure N112 und/oder der ECL1-Domäne am Ionen-Translokationsweg, sowie das enge Zusammenspiel zwischen dem Retinal und seinem Gegenion D116. Gleichzeitig bot diese IR-Studie einen der ersten mechanistischen Einblicke in den Protonenpump-Photozyklus in KR2, der deutlich weniger erforscht war. In Zusammenarbeit mit dem Arbeitskreis Glaubitz wurden die strukturellen Veränderungen des Chromophors und seiner Umgebung während der verschiedenen Photointermediate mittels DNP-verstärkter Festkörper-NMR und optischer Spektroskopie näher untersucht. Hier trugen zeitaufgelöste IR-Messungen in der HOOP (engl. hydrogen out of plane)-Moden-Region dazu bei, die dynamischen Veränderungen der Chromophorkonfiguration und -Verdrillung zu verfolgen. Es konnte gezeigt werden, dass Retinal im O-Intermediat tatsächlich zu seiner all-trans-Konfiguration reisomerisiert wird, aber im Vergleich zu seiner Dunkelzustandskonfiguration deutlich stärker verdreht vorliegt.
Dies wurde auch durch die Ergebnisse im nahen UV-Bereich bestätigt, welcher bei der Charakterisierung von mikrobiellen Rhodopsinen oft ausgelassen wird. Die neu gefundene Signatur erwies sich als SBS (engl. second bright state) der 13-cis-Konfiguration des Retinals, die mit der Bildung des O-Intermediats in KR2 verschwindet. Neben der offensichtlichen Verwendung als spektraler Marker wurde der SBS-Übergang auch bezüglich seiner Anwendbarkeit für optische Kontrollexperimente untersucht. Ähnlich wie beim BLQ (engl. blue light quenching)-Effekt war es möglich, den KR2-Dunkelzustand durch Anwendung von fs-Pulsen im nahen UV - ausgehend von einem photostationären Zustand - zu regenerieren. Durch Variation der Probenbedingungen war es möglich, gezielt K (pH~5) oder M (pH~9) anzusteuern, was sich auch in unterschiedlichen Deaktivierungs-Dynamiken äußerte. Diese Ergebnisse können zusammen mit dem hier vorgeschlagenen experimentellen Konzept als Grundlage für komplexere Multiphotonen-Sequenzen im Zusammenhang optogenetischer Fragestellungen verwendet werden.
Im Gegensatz zu den recht großen und komplexen Photorezeptoren bieten unter anderem PPGs einen feineren Weg, um biochemische Reaktionen gezielt zu steuern und auszulesen. In diesem Zusammenhang sind zwei Eigenschaften von großer Bedeutung: Einerseits die Fähigkeit der PPG, Photonen bestimmter Wellenlängen zu absorbieren, und andererseits die Effizienz der gewünschten photochemischen Reaktion. Der letztgenannte Aspekt wurde unter der Hypothese untersucht, dass die Verringerung der konkurrierenden Deaktivierungskanäle in PPGs zu einer höheren Quantenffizienz der Freisetzung führt. Dies wurde an DEACM-basierten Modellverbindungen getestet, die systematisch modifiziert wurden, um verschiedene Deaktivierungsprozesse des angeregten Zustands zu identifizieren. Durch das Hinzufügen eines zusätzlichen sechsgliedrigen Rings wurde die Freisetzungsausbeute im Vergleich zu DEACM um das 2- bis 3-fache erhöht. Dies konnte durch eine weitere Planarisierung des Systems mit einer zusätzlichen Doppelbindung an der C6-Position sogar noch weiter verbessert werden (bis zu einem Faktor von 5-6). Die Anregung des Cumarin-Rückgrats führt zu einem lokal-angeregten Zustand, der sich im Gleichgewicht mit einem Ladungstransferzustand befindet. In Abhängigkeit der lokalen Umgebung, die vor allem durch die Protizität und Polarität des Lösungsmittels bestimmt wird, wird der Ladungstransfercharakter eher stabilisiert oder gar destabilisiert. Die Ladungsverschiebung führt auch zu einer Abschwächung der spaltbaren C-C-Bindung, die eine Voraussetzung für die Freisetzungsreaktion ist. Darüber hinaus wurde gezeigt, dass der mit der Freisetzungsreaktion verbundene zusätzliche Zerfallskanal zu einer mehr als 2-fachen Verringerung der Lebensdauer des angeregten Zustands in den funktionalisierten PPGs führt. Diese Eigenschaft ist ein vielversprechender photophysikalischer Indikator für die Freisetzung der Abgangsgruppe, der durch spektroskopische oder - mit zusätzlicher räumlicher Auflösung - auch durch mikroskopische Techniken wie in der Fluoreszenzlebensdauer-Mikroskopie ausgelesen werden könnte...
Die Arbeit befasst sich mit einer Vereinfachung des von Devroye (1999) geprägten Begriffs der random split trees und verallgemeinert diesen im Sinne von Janson (2019) auf unbeschränkten Verzweigungsgrad. Diese Verallgemeinerung deckt auch preferential attachment trees mit linearen Gewichten ab, wofür ein Beweis von Janson (2019) aufbereitet wird. Zusätzlich bleiben die von Devroye (1999) nachgewiesenen Eigenschaften über die Tiefe der hinzugefügten Knoten erhalten.
Die vorliegende Arbeit untersucht den Übergang von Mitarbeitenden zu Führungskräften. Der Fokus der Arbeit ist nicht die Konzeption der Führungskräfteentwicklung aus der Perspektive der Unternehmen, sondern die Untersuchung der Entstehung und Gestaltung dieses Übergangs durch die Mitarbeitenden innerhalb ihrer beruflichen Rahmenbedingungen. Die Ergebnisse dieser Arbeit zeigen, dass sich in der Gestaltung dieses Übergangs von Mitarbeitenden zur Führungskräften individuelle und organisatorische Aspekte verbinden – bzw. diese durch das Subjekt in eine Verbindung gebracht werden. Durch leitfadengestützte Interviews mit (berufs-) biografischem Schwerpunkt ließen sich die individuellen, wie auch die sozialen und organisatorischen Aspekte der Gestaltung des Übergangs von Mitarbeitenden zur Führungskräften rekonstruieren. Die Stichprobe dieser Arbeit umfasst neun Interviews aus unterschiedlichsten Branchen und beinhaltet sowohl öffentliche als auch privatwirtschaftliche Arbeitgeber. Die dadurch gewonnenen Erkenntnisse ermöglichen Einsichten in die Formen und Bedingungen der Gestaltung dieses Übergangs und können auch eine Grundlage für (organisations-)pädagogische Konzepte zur Unterstützung der Mitarbeitenden bei diesem Übergang bilden.
Die Magisterarbeit in ein Beitrag zur Weiterentwicklung neuzeitlicher Subjekt- und Emanzipationstheorien. Im ersten Schritt werden die Grundkonzepte aus Baruch de Spinozas Ethik dargestellt und ihre Rolle fuer die Ideologietheorie Louis Althussers analysiert. Auf Grundlage dieser Analyse werden Grenzen der Subjekt- und Ideologietheorie Altussers aufgezeigt und der Versuch unternommen sie unter Zugriff auf spinozistische Begigriffe zu ueberwinden. Eine zentrale Rolle spielt hierbei die Perspektive auf den Menschen als körperliches, emotionales und begehrendes Wesen.
First-principles modeling techniques offer the ability to simulate a wide range of systems under different physical conditions, such as temperature, pressure, and composition, without relying on empirical knowledge. Density functional theory (DFT), a quantum mechanical method, has become an exceptionally successful framework for materials science modeling. Employing DFT makes it possible to gain valuable insights into the fundamental state of a system, enabling the reliable determination of equilibrium crystal structures. Over time, DFT has become an essential tool that can be incorporated into various schemes for predicting the properties of a material related to its structure, insulating/metallic behavior, magnetism, and optics. DFT is regularly applied in numerous fields, spanning from fundamental subjects in condensed matter physics to the study of large-scale phenomena in geosciences. In the latter, the effectiveness of DFT stems from its ability to simulate the properties found on the Earth, other planets, and meteorites, which may pose challenges for their direct study or laboratory investigation.
In this thesis, a comprehensive examination of a family of monosulfides and a perovskite heterostructure was conducted. These materials are relevant for their potential applications in technology, energy harvesting, and in the case of monosulfides, their speculated abundance on the planet Mercury.
Firstly, a DFT approach was used to analyze two non-magnetic monosulfides, CaS and MgS. We determined their structural properties and then focused on the modeling of their reflectivity in the infrared region. The calculation of the reflectivity considered both harmonic and anharmonic contributions. In the harmonic limit, the non-analytic correction was employed to accurately determine the LO/TO splitting, which is necessary to delimit the retstrahlend band, that is, the maximum of the reflectivity. The anharmonic effects given by up to three-phonon and isotopic scatterings, which were included using perturbation theory, primarily smeared the reflectivity spectra edges in the high-wave region.
Secondly, four polymorphs of MnS were studied using a combination of first-principles methods to simulate their antiferromagnetic (AFM) and paramagnetic (PM) states. The integration of DFT+$U$ with special quasirandom structures (SQS) supercells, and occupation matrix control techniques was crucial for achieving convergence, structural optimization accuracy, and obtaining finite energy band gaps and local magnetic moments in the PM phases. The addition of the Hubbard $U$ correction was necessary to treat the highly-correlated Mn $d$-electrons. The success of our approach was clear based on our electronic structure predictions for the PM rock-salt B1-MnS polymorph. Experimentally this phase has been observed to be an insulator, but multiple \emph{ab initio} works resulted previously in metallic behavior. Our computations, on the other hand, predicted insulating and magnetic properties that compare well with available measurements. Additionally, the pressure-field stability of the four MnS polymorphs was studied. In the case of the PM phases, B1-MnS was identified to be the most stable up to about 21 GPa, then transforming into the B31-MnS polymorph. This finding was in close agreement with high-pressure experiments reporting a similar phase transformation. The optical properties of B1-, B4-, and B31-MnS were also simulated. The SQS technique was used to obtain soft-mode-free phonon band structures within the harmonic approximation. Then, the anharmonic effects were included, and the reflectivity was calculated for B1-MnS and B4-MnS. In both cases, a good agreement for the LO/TO splitting was achieved in comparison to experimental results.
Lastly, the oxygen-deficient heterostructure of LaAlO$_{3-\delta}$ /SrTiO$_{3-\delta}$ was investigated also employing DFT+$U$, with a particular emphasis on the potential impact of vacancy clustering at the interface. Six distinct configurations of pairs of vacancies were studied and their energies were compared to find the most stable one. The orbital reconstruction of Ti orbitals was also examined based on their location with respect to the vacancies and the local magnetic moments were calculated. The final results showed that linearly arranged vacancies located opposite to Ti ions give the most energetically stable configuration.
Die vorliegende Dissertation befasst sich mit Flow-Zuständen beim Lesen fiktiver Texte. Das 1975 von Mihaly Csikszentmihalyi vorgestellte Konzept des Flow bezieht sich auf das völlige Aufgehen in einer optimal herausfordernden Tätigkeit, das mit Absorption, Verarbeitungsflüssigkeit und intrinsische Freude einhergeht. Bislang wurde Flow zumeist im Kontext motorischer und leistungsorientierter Aktivitäten empirisch untersucht und in erster Linie theoretisch mit Lesefreude in Verbindung gebracht. Ziel der drei Studien, die diese Dissertation umfasst, war es daher einerseits, Flow beim Lesen erstmals anhand größerer Leser-Stichproben und mithilfe von psychometrischen Gütekriterien genügenden Messinstrumenten nachzuweisen. Andererseits sollte Flow im Rahmen eines Modells für positives Leseerleben mit anderen in der Leseforschung diskutierten Konzepten in Verbindung gebracht und im Hinblick auf potenzielle psychophysiologische Korrelate untersucht werden.
In der ersten Studie wurde eine in der allgemeinen Flow-Forschung verbreitete Kurz-Skala an den Lesekontext adaptiert und anhand einer 229 Leser umfassenden Stichprobe psychometrisch getestet. Hierzu wurden die Teilnehmer im Rahmen einer Online-Studie gebeten, nach 20-minütigem Lesen in einem selbstgewählten Roman Fragebögen zu ihrem Leseerleben auszufüllen. Zufriedenstellende Reliabilitätskoeffizienten, positive Korrelationen mit konvergenten Maßen, die faktoranalytische Unterscheidbarkeit zu diskriminanten Maßen und die erwartete Assoziation mit einem Flow-Kriterium bestätigten die Güte der Flow-Skala. Eine Explorative Faktorenanalyse ergab jedoch, dass fast alle Items auf dem Faktor Absorption luden. Zudem ließ die zweifakorielle Skalenstruktur keine abschließende Aussage zur Legitimierung eines globalen Flow-Scores zu. Daher wurde in der zweiten Studie auf Basis der ersten Skala und der aus der Theorie bekannten Flow-Komponenten ein umfassenderer lesespezifischer Flow-Fragebogen entwickelt. Dessen Reliabilität und Validität konnte anhand einer Online-Studie mit 373 Teilnehmern, in deren Rahmen ein Kapitel aus Homers Odyssee gelesen wurde, bestätigt werden. Neben Hinweisen zur konvergenten und diskriminanten Konstrukt- und zur Kriteriumsvalidität stützten die Ergebnisse einer Konfirmatorischen Faktorenanalyse eine theoretisch angemessene Skalenstruktur, mit den einzelnen Komponenten, mit Absorption, Verarbeitungsflüssigkeit und intrinsischer Freude als Subdimensionen und mit Flow als übergeordnetem Faktor. Mittels eines Strukturgleichungsmodells konnte zudem demonstriert werden, dass der auf Basis dieses Fragebogens gemessene Flow eine zentrale Rolle beim Leseerleben einnehmen kann. So wurde Flow als Mediator für andere, ebenfalls erhobene Erlebnisformen beim Lesen wie etwa Identifikation oder Spannung bestätigt. Von diesen Konzepten klärte Flow den größten Anteil an Varianz in Lesefreude und Textverständnis auf, die als Outcomes von positivem Leseerleben modelliert wurden. Da Flow gegenüber anderen Konzepten der Leseforschung den Vorteil hat, die Ableitung experimenteller Paradigmen und psychophysiologischer Hypothesen zu ermöglichen, wurden in der dritten Studie über die Manipulation des stilistischen Herausforderungsgrades eines weiteren Odyssee-Kapitels unterschiedliche Lese-Bedingungen hergestellt und kardiovaskuläre Daten gemessen. Es zeigten sich zwar keine signifikanten Gruppenunterschiede im Flow-Erleben, jedoch Interaktionen zwischen der Lesebedingung und kardiovaskulären Indikatoren bei der Vorhersage von Flow. So scheinen parasympathische Dominanz und ein entsprechender innerer Entspannungszustand, indiziert durch eine geringe Herzrate und hohe Herzratenvariabilität, Flow beim Lesen zu begünstigen, wenn der Text stilistisch anspruchsvoll ist. Es fanden sich hingegen keine Hinweise dafür, dass Flow-Erleben die Herzaktivität von Lesern verändert oder sich durch sie objektiv erfassen lässt.
Insgesamt sprechen die Ergebnisse dieses Forschungsprojektes somit für das Auf-treten von Flow beim Lesen sowie für dessen zentrale Rolle bei positiven Leseerlebnissen. Außerdem zeigen sie das Potenzial des Flow-Konzeptes für die Leseforschung auf, insbesondere hinsichtlich psychophysiologischer Experimentalstudien.
