Motivated by the on-going discussion on the nature of magnetism in the quantum Ising chain CoNb2O6, we present a first-principles-based analysis of its exchange interactions by applying an \textit{ab initio} approach with additional modelling that accounts for various drawbacks of a purely density functional theory ansatz. With this method we are able to extract and understand the origin of the magnetic couplings under inclusion of all symmetry-allowed terms, and to resolve the conflicting model descriptions in CoNb2O6. We find that the twisted Kitaev chain and the transverse-field ferromagnetic Ising chain views are mutually compatible, although additional off-diagonal exchanges are necessary to provide a complete picture. We show that the dominant exchange interaction is a ligand-centered exchange process - involving the eg electrons -, which is rendered anisotropic by the low-symmetry crystal fields environments in CoNb2O6, giving rise to the dominant Ising exchange, while the smaller bond-dependent anisotropies are found to originate from d−d kinetic exchange processes involving the t2g electrons. We demonstrate the validity of our approach by comparing the predictions of the obtained low-energy model to measured THz and inelastic neutron scattering spectra.
Raumkonstruktionen, die über Handlung und Bedeutungszuschreibungen im Kontext sozialer Medien entstehen, sind ein Fallbeispiel dafür, wie vor dem Hintergrund von Digitalität Fachinhalte re-innoviert werden müssen. Mit dem Ziel eines Beitrags zur Konzeption professionellen Lehrkräftewissens im Kontext von Digitalität werden, ausgehend von der Fragestellung, welche professionellen Fähigkeiten Lehrkräfte benötigen, um Raumkonstruktionen im Unterricht zu thematisieren, in dieser kumulativen Dissertation Raumkonstruktionen aus normativer und empirischer Perspektive als exemplarischer geographischer Fachinhalt adressiert.
Als theoretischer Rahmen dient dabei das TPACK Modell von Mishra & Koehler (2006), das professionelles Lehrkräftewissen in die Bereiche fachliches, pädagogisches und technologisches Wissen sowie deren Überschneidungsbereiche einteilt. Zunächst erfolgt eine Anwendung des Modells als Reflexionsperspektive auf Fachinhalte zur Erzeugung normativer Fähigkeitsbeschreibungen. Diese Fähigkeitsbeschreibungen fließen in die Entwicklung eines TPACK-Selbsteinschätzungsfragebogens für Lehramtsstudierende der Geographie ein. Im Rahmen der damit durchgeführten Studie (n= 364) zeigen sich auf deskriptiver Ebene vergleichsweise niedrige Selbsteinschätzungen der Bereiche inhaltlichen und fachdidaktischen Wissens. Durch die Anwendung einer konfirmatorischen Faktorenanalyse kann das TPACK Modell als zufriedenstellend für die Beschreibung der Daten identifiziert werden. Auffällig ist allerdings die niedrige Korrelation des Konstrukts technologischen Wissens mit den angrenzenden Wissensbereichen. In Bezug auf die Selbsteinschätzungen im phasen- und kontextübergreifenden Vergleich lässt sich, ausgehend von linearen Regressionsanalysen, eine tendenzielle Zunahme der selbsteingeschätzten Fähigkeiten entlang der Fachsemesterzahl ermitteln. Mittels Zweistichproben-t-Tests können außerdem höhere Selbsteinschätzungen der pädagogischen Wissensbereiche durch Studierende, die als Vertretungslehrkräfte tätig sind, festgestellt werden.
In Bezug auf die zur Thematisierung von Raumkonstruktionen im Unterricht benötigten Fähigkeiten ist zunächst die Relevanz der Förderung der Integration fachlichen, pädagogischen und fachdidaktischen Wissens hervorzuheben. Die Studie gibt darüber hinaus Hinweise auf eine niedrigere Bedeutung technologischen Wissens im Hinblick auf soziale Medien als Beispiele alltäglicher Technologien. Vor dem Hintergrund des positiven Effekts von Selbstwirksamkeitsprozessen bieten die Ergebnisse Implikationen für eine Diskussion von praktischen Erfahrungen als Aspekt professionellen Lehrkräftewissens im Sinne der Förderung einer kritisch-reflexiven Auseinandersetzung mit der Tätigkeit als Vertretungslehrkraft aus fachdidaktischer Perspektive. Insgesamt leistet die kumulative Dissertation einen Beitrag zum Diskurs um fachliche geographische Bildung im Kontext von Digitalität und zur Konzeption des professionellen Lehrkräftewissens vor diesem Hintergrund.
Der Dialog ist für die einen das Versprechen gelingender Kommunikation, für die anderen ein überholtes Ideal. Marten Weise zeigt in einer interdisziplinär angelegten Studie, dass sich die Lücke zwischen Lobpreisungen und Abgesängen schließen lässt. Er setzt bei der Unmöglichkeit des Denkens "nach der Shoah" an und erkundet in exemplarischen Untersuchungen der europäischen Literatur-, Theater- und Theoriegeschichte die Spannungen und Widersprüche im Verhältnis zum "Anderen", ohne die der Dialog nicht zu greifen ist. So macht er zwischenmenschliche, soziale und politische Vorgänge als prinzipiell unabschließbares Sprachgeschehen fassbar und eröffnet einen Spielraum für die Aushandlung und das Aushalten von Dissens und Differenz.
The knowledge of the material budget with a high precision is fundamental for measurements of direct photon production using the photon conversion method due to its direct impact on the total systematic uncertainty. Moreover, it influences many aspects of the charged-particle reconstruction performance. In this article, two procedures to determine data-driven corrections to the material-budget description in ALICE simulation software are developed. One is based on the precise knowledge of the gas composition in the Time Projection Chamber. The other is based on the robustness of the ratio between the produced number of photons and charged particles, to a large extent due to the approximate isospin symmetry in the number of produced neutral and charged pions. Both methods are applied to ALICE data allowing for a reduction of the overall material budget systematic uncertainty from 4.5% down to 2.5%. Using these methods, a locally correct material budget is also achieved. The two proposed methods are generic and can be applied to any experiment in a similar fashion.
Memory consolidation tends to be less robust in childhood than adulthood. However, little is known about the corresponding functional differences in the developing brain that may underlie age-related differences in retention of memories over time. This study examined system-level memory consolidation of object-scene associations after learning (immediate delay), one night of sleep (short delay), as well as two weeks (long delay) in 5-to-7-year-old children (n = 49) and in young adults (n = 39), as a reference group with mature consolidation systems. Particularly, we characterized how functional neural activation and reinstatement of neural patterns change over time, assessed by functional magnetic resonance imaging combined with representational similarity analysis (RSA). Our results showed that memory consolidation in children was less robust and strong (i.e., more forgetting) compared to young adults. Contrasting correctly retained remote versus recent memories across time delay, children showed less upregulation in posterior parahippocampal gyrus, lateral occipital cortex, and cerebellum than adults. In addition, both children and adults showed decrease in scene-specific neural reinstatement over time, indicating time-related decay of detailed differentiated memories. At the same time, we observed more generic gist-like neural reinstatement in medial-temporal and prefrontal brain regions uniquely in children, indicating qualitative difference in memory trace in children. Taken together, 5-to-7-year-old children, compared to young adults, show less robust memory consolidation, possibly due to difficulties in engaging in differentiated neural reinstatement in neocortical mnemonic regions during retrieval of remote memories, coupled with relying more on gist-like generic neural reinstatement.
Long- and short-range correlations for pairs of charged particles are studied via two-particle angular correlations in pp collisions at √sNN = 13 TeV and p–Pb collisions at √s = 5.02 TeV. The correlation functions are measured as a function of relative azimuthal angle ∆φ and pseudorapidity separation ∆η for pairs of primary charged particles within the pseudorapidity interval |η| < 0.9 and the transverse-momentum interval 1 < pT < 4 GeV/c. Flow coefficients are extracted for the long-range correlations (1.6 < |∆η| < 1.8) in various high-multiplicity event classes using the low-multiplicity template fit method. The method is used to subtract the enhanced yield of away-side jet fragments in high-multiplicity events. These results show decreasing flow signals toward lower multiplicity events. Furthermore, the flow coefficients for events with hard probes, such as jets or leading particles, do not exhibit any significant changes compared to those obtained from high-multiplicity events without any specific event selection criteria. The results are compared with hydrodynamic-model calculations, and it is found that a better understanding of the initial conditions is necessary to describe the results, particularly for low-multiplicity events.
The production cross section of inclusive isolated photons has been measured by the ALICE experiment at the CERN LHC in pp collisions at centre-of-momentum energy of s√=13 TeV collected during the LHC Run 2 data-taking period. The measurement is performed by combining the measurements of the electromagnetic calorimeter EMCal and the central tracking detectors ITS and TPC, covering a pseudorapidity range of |ηγ|<0.67 and a transverse momentum range of 7<pγT<200 GeV/c. The result extends to lower pγT and xγT=2pγT/s√ ranges, the lowest xγT of any isolated photon measurements to date, extending significantly those measured by the ATLAS and CMS experiments towards lower pγT at the same collision energy with a small overlap between the measurements. The measurement is compared with next-to-leading order perturbative QCD calculations and the results from the ATLAS and CMS experiments as well as with measurements at other collision energies. The measurement and theory prediction are in agreement with each other within the experimental and theoretical uncertainties.
Inorganic phosphate is one of the most abundant and essential nutrients in living organisms. It plays an indispensable role in energy metabolism and serves as a building block for major cellular components such as the backbones of DNA and RNA, headgroups of phospholipids and in posttranslational modifcations of many proteins. Disturbances in cellular phosphate homeostasis have a detrimental effect on the viability of cells. There- fore, both the import and export of phosphate is strictly regulated in eukaryotic cells. In the eukaryotic model organism Saccharomyces cerevisiae, the uptake of phosphate is carried out either by transporters with high affinity or by transporters with low affinity, depending on the cytosolic phosphate concentration. While structures are available for homologues of the high-affinity transporters, no structures of low-affinity transporters have been solved so far. Interestingly, only the low-affinity transporters have a regulatory SPX domain, which is found in various proteins involved in phosphate homeostasis.
In this work, structures of Pho90 from Saccharomyces cerevisiae, a low-affinity phosphate transporter, were solved by cryo-EM, providing insights into its transport mechanism. The dimeric structure resembles the structures of proteins of the divalent anion symporter superfamily (DASS) and of mammalian transporters of the solute carrier 13 (SLC13) family. The transmembrane domain of each protomer consists of 13 helical elements and can be subdivided into scaffold and transport domains. The structure of ScPho90 in the presence of phosphate shows the phosphate binding site within the transporter domain in an outward-open conformation with a bound phosphate ion and two sodium ions. In the absence of phosphate, an asymmetric dimer structure was determined, with one protomer adopting an inward-open conformation. While the dimer contact and the scaffold domain are identical in both conformations, the transport domain is rotated by about 30° and shifted by 11 Å towards the cytoplasmic side, leading to the accessibility of the binding pocket from the cytoplasm. Based on these findings and by comparison with known structures, a phosphate transport mechanism is proposed in the present work that involves substrate binding on the extracellular side, conformational change by a rigid-body motion of the transport domain, in an "elevator-like" motion, and substrate release into the cytoplasm. The regulatory SPX domain is not well resolved in the ScPho90 structures, so that no direct conclusions were drawn about its regulatory mechanism. The findings provide new insights into the function and mechanism of eukaryotic low-affinity phosphate transporters.
While eukaryotic cells express various phosphate import proteins, most eukaryotes have only a single highly conserved and essential phosphate exporter. These exporters show no sequence homology to other transporters of known structure, but also possess a regulatory SPX domain. In this work, the structural basis for eukaryotic phosphate export is investigated by elucidating the structures of the homologous phosphate exporters Syg1 from Saccharomyces cerevisiae and Xpr1 from Homo sapiens, using cryo-EM. The structures of ScSyg1 and HsXpr1 show a conserved homodimeric structure and the transmembrane part of each protomer consists of 10 TM helices. Helix TM1 establishes the dimer contact by means of a glycine zipper motif, which is a known oligomerization motif. Helices TM2-5 form a hydrophobic pocket that has density for a lipid molecule. Whether the lipid binding into the hydrophobic pocket has an allosteric effect on the phosphate export activity or only serves protein stabilization is not known. Helices TM5-10 form a six-helix bundle, which constitutes a putative phosphate translocation pathway in its center. This bundle is formed by the protein sequence annotated as EXS domain.
The respective phosphate translocation pathways of ScSyg1 and HsXpr1 show structural differences. While the translocation pathway in HsXpr1 is accessible from the cytoplasm, in ScSyg1 it is closed by a large loop of the SPX domain. Interestingly, this loop is not conserved in higher eukaryotes and is therefore not present in HsXpr1. Another difference are distinct conformations of helix TM9. In ScSyg1, TM9 adopts a kinked conformation, which results in the translocation pathway being open to the extracellular side. In contrast, TM9 adopts a straight conformation in HsXpr1, resulting in the placement of a highly conserved tryptophane residue in the middle of the translocation pathway. As a result, the translocation pathway in HsXpr1 is closed to the extracellular side.
The e+e−→D+sDs1(2536)− and e+e−→D+sD∗s2(2573)− processes are studied using data samples collected with the BESIII detector at center-of-mass energies from 4.530 to 4.946~GeV. The absolute branching fractions of Ds1(2536)−→D¯∗0K− and D∗s2(2573)−→D¯0K− are measured for the first time to be (35.9±4.8±3.5)% and (37.4±3.1±4.6)%, respectively. The measurements are in tension with predictions based on the assumption that the Ds1(2536) and D∗s2(2573) are dominated by a bare cs¯ component. The e+e−→D+sDs1(2536)− and e+e−→D+sD∗s2(2573)− cross sections are measured, and a resonant structure at around 4.6~GeV with a width of 50~MeV is observed for the first time with a statistical significance of 15σ in the e+e−→D+sD∗s2(2573)− process. It could be the Y(4626) found by the Belle collaboration in the D+sDs1(2536)− final state, since they have similar masses and widths. There is also evidence for a structure at around 4.75~GeV in both processes.
Using a sample of 448.1×106 ψ(2S) events collected with the BESIII detector, we perform a study of the decay J/ψ→K+K− via ψ(2S)→π+π−J/ψ.
The branching fraction of J/ψ→K+K− is determined to be BK+K−=(3.072±0.023(stat.)±0.050(syst.))×10−4, which is consistent with previous measurements but with significantly improved precision.
In dieser Dissertation werden die Erfahrungen mit verschiedenen Präparationsmethoden für CH-Kavitäten beschrieben, um die Leistung der Kavitäten nach der Herstellung weiter zu steigern. Die Leistung wird anhand von zwei wichtigen HF-Parametern bewertet:
dem elektrischen Feld Ea und der intrinsischen Güte Q0. Im Gegensatz zu normalleitenden (NC) Kavitäten kann die intrinsische Güte von supraleitenden (SC) Kavitäten mit zunehmendem elektrischem Feld erheblich variieren. Das optimale Ergebnis für die Kavitätenpräparation ist die Erhöhung des maximalen elektrischen Feldes unter Beibehaltung eines höheren Q0 über die gesamte Feldspanne. Da Q0 umgekehrt proportional zu den Kavitätsverlusten ist, reduziert eine Erhöhung des Qualitätsfaktors die Kryoverluste für den Betrieb bei gegebenem Feldniveau. Die Entwicklung der Kavitätenperformanz im Verlauf dieser Arbeit dargestellt.
Die meisten SC-Kavitäten sind elliptische Strukturen, welche bei hoher Geschwindigkeit und Tastrate angewendet werden. Die Präparationsmethoden wurden daher überwiegend auf diese Strukturen angewandt und optimiert. Diese Arbeit konzentriert sich auf die Umsetzung der zuverlässigsten und vielversprechendsten Oberflächenbehandlungen mittels des ersten vom IAP entwickelten SC 360MHz CH-Prototyps. Diese Kavität wies nach 11 Jahren Lagerung eine verminderte Leistung auf, welche mit Röntgenstrahlung bei bereits niedrigen elektrischen Feldern einherging. Dies deutet auf eine unbeabsichtigte Belüftung mit normaler Luft hin, durch die Partikel eingeführt wurden, die als verstärkende Quellen von Elektronen fungierten. Außerdem musste der Leistungskoppler aufgrund einer starken Überkopplung neu ausgelegt werden.
Die Kavität wurde für 48 Stunden bei 120◦ C mittels Heizbändern in der Experimentierhalle des IAP’s ausgeheizt, was zu einer Verbesserung des Qualitätsfaktors bei niedrigen Werten und zu einer Verkürzung der für die Konditionierung von Multipacting-Barrieren erforderlichen Zeit führte. Allerdings wurde durch diese Behandlung das maximale erreichbare elektrische Feld weiter verringert. Die Verbesserung der Güte ist auf das Ausgasen der Kohlenwasserstoffe während des Backvorgangs zurückzuführen. Die negative Auswirkung auf das maximale elektrische Feld ist weniger auf das Backen selbst zurückzuführen als auf den Transport der Kavität und die verwendeten Vakuumkomponenten, die in der Versuchshalle gelagert sind.
Die beobachtete Leistungseinschränkung lässt sich hauptsächlich durch Partikel im Inneren des Resonators erklären, da Feldemission bei niedrigen Feldstärken auftrat. Eine Hochdruckspülung mit ultrareinem Wasser (HPR) ist das Standardverfahren, um nach Behandlungen, bei denen das Risiko einer Oberflächenkontamination besteht, eine hohe Reinheit der inneren Oberflächen zu erreichen. Die HPR wurde in Zusammenarbeit mit dem Helmholtz-Institut-Mainz und der Gesellschaft für Schwerionenforschung geplant und durchgeführt. Der Resonator zeigte bereits während der Messung der Q-E-Kurve eine Zunahme der transmittierten Leistung bei konstanter Vorwärtsleistung, was vor der HPR nicht der Fall war. Bei der CW-HF-Konditionierung zeigte die Kavität den höchsten Gradienten bei einem deutlich schwächeren Q-Abfall bei hohen Feldstärken.
Sowohl bei der Messung von 2008 als auch bei der beschriebenen Messung wurde die Kavität mit einer HPR-Behandlung fertiggestellt, aber für die HPR-Behandlung bei HIM in Mainz wurden einige Anpassungen vorbereitet. Der CH Prototyp verfügt über keine zusätzlichen Spülports und wurde daher mit zwei verschiedenen Düsen mit unterschiedlichen Sprühwinkeln gespült, um die erreichbaren inneren Resonatorflächen zu maximieren. Die Verwendung mehrerer Sprühwinkel könnte auch für CH-Kavitäten mit Spülöffnungen von Vorteil sein und sollte für zukünftige HPR-Anwendungen in Betracht gezogen werden.
Die Heliumbehandlung wurde am CH-Prototyp 2,5 Stunden lang durchgeführt und lieferte vielversprechende Ergebnisse in Bezug auf die Güte und die Gradientenoptimierung.
Während dieses Prozesses wurde die emittierte Röntgenstrahlung in Richtung am Arbeitsplatzs gemessen und zeigte starke zeitabhängige Fluktuationen. Dies deutete auf die Beseitigung von Partikeln hin und wurde anschließend durch einen Anstieg des elektrischen Feldes von 8,4 auf 8,7 MV/m bestätigt. Eine unerwartete Auswirkung wurde bei der Q-Steigung im mittleren bis hohen Feld festgestellt, bei der der Qualitätsfaktor im Vergleich zum HF-konditionierten Fall eine Erhöhung von 5% oberhalb von 2MV/m aufwies. Dieser systematische Anstieg wurde für diesen Beschleuniger vor der Behandlung bisher nicht beobachtet. Stickstoffgedopte Kavitäten zeigen ein ähnliches Verhalten, bei dem Wechselwirkungen innerhalb der Oxidschicht mit Änderungen der Qualitätsfaktoren korreliert sind. Da Helium ein nicht reaktives Element ist, sind mögliche Erklärungen für diesen Effekt der Sputterprozess und die Einlagerung von Helium innerhalb der Oberfläche. Eine Serie von Heliumbehandlungen ist geplant, um ein optimiertes und sicheres Rezept für CH-Kavitäten zu finden. Die Q-E-Messung nach der Abkühlung und vor der Behandlung wird auch zeigen, ob der Leistungsgewinn durch ein Aufwärmen auf Raumtemperatur beeinträchtigt wird.
Die in dieser Arbeit skizzierte Behandlungssequenz wird für CH-Kavitäten dringend empfohlen. Das Ausheizen hat sich bei der Verringerung des Multipactings and der Güteabnahme bei hohen Feldern als wirksam erwiesen und bleibt von der anschließenden HPR unbeeinflusst. In dieser Arbeit wurden keine negativen Auswirkungen der HPR auf das Multipactingverhalten festgestellt. Anschließend wird eine CW-HF-Konditionierung durchgeführt, bis keine weitere Leistungszunahme der Kavität mehr zu verzeichnen ist.
Wenn die Kavität immer noch durch Feldemission begrenzt ist, sollte eine Wiederholung der HPR-Behandlung in Betracht gezogen werden, da bei sorgfältiger Durchführung der HPR keine der bisherig gefertigten CH-Kavitäten hierdurch begrenzt war. Es ist auch anzumerken, dass die Heliumbehandlung nur an der 360MHz CH-Kavität durchgeführt wurde, als diese eine geringe Strahlung durch Feldemission aufwies. Das Risiko des Heliumprocessing an CH-Kavitäten unter starker Feldemission ist unbekannt. Es ist zu erwarten, dass die Elektronenströme und damit die Ionenbeschusslawinen zunehmen und ein größeres Risiko für die Beschädigung von der Komponenten darstellen. Nach dem derzeitigen Kenntnisstand sollte die Heliumbehandlung nur für gut vorbereitete Kavitäten mit minimaler Feldemission in Betracht gezogen werden.
An independent Taiwanese lineage of powdery mildew on the endemic host species Koelreuteria henryi

Background: Powdery mildews (Erysiphaceae, Ascomycota) are common plant disease agents and also cause stress for forest and fruit trees worldwide as well as in Taiwan. The powdery mildew Erysiphe bulbouncinula on Koelreuteria host trees was considered an endemic species in China. While in China the host was K. paniculata and only the teleomorph stage found, the anamorph and the teleomorph were both recorded for the host in Taiwan, K. henryi. We aimed to clarify the relationship of the powdery mildews recorded under E. bulbouncinula with an apparently disjunct distribution.
Results: Specimens of powdery mildew on K. henryi from Taiwan were characterized based on the anamorph morphology and DNA sequences. They revealed a new record of Sawadaea koelreuteriae for this host species and Taiwan and a new species of Erysiphe, E. formosana, sister to E. bulbouncinula from China.
Conclusions: In Erysiphe on Koelreuteria hosts, speciation of plant parasitic fungi seems to be correlated with disjunct host and geographic distribution possibly shaped by extinction of potential host species which are known only as fossils. Two of the three extant East Asian species of Koelreuteria are now known as hosts of specific Erysiphe species. We may predict a further not yet discovered Erysiphe species on the third East Asian species, K. bipinnata, in South and Southwest China. In the speciation in Sawadaea, the extinction events in Koelreuteria can be excluded from being involved.
Das partizipative Forschungsprojekt "Allein aber vernetzt? Digitale (Un)gleichheiten und soziale Netzwerke bei alleinlebenden Menschen" beschäftigte sich von 2021-2024 mit Alleinlebenden im Alter, ihren Beziehungsnetzwerken und der Frage welche Rolle digitale Geräte und die Digitalisierung aller Lebensbereiche in diesem Zusammenhang spielen. Durch die Zusammenarbeit mit einer Gruppe von Ko-Forschenden in Frankfurt und Egelsbach stand die Perspektive der Gruppe im Mittelpunkt.
Im vorliegenden White Paper wird das Projekt sowie zentrale Ergebnisse und die erarbeiteten Handlungsempfehlungen der deutschen Teilstudie vorgestellt. Diese wurde vom BMBF gefördert und ist Teil des europäischen Verbundprojektes "EQualCare - Alone but connected? Digital (in)equalities in care work and generational relationships among older people living alone" der Joint Programming Initiative (JPI) “More Years Better Lives”.

Based on 4.5 fb−1 of e+e− collision data accumulated at center-of-mass energies between 4599.53 MeV and 4698.82 MeV with the BESIII detector, the decay Λ+c→nK0Sπ+π0 is observed for the first time with a significance of 9.2σ. The branching fraction is measured to be (0.85±0.13±0.03)%, where the first uncertainty is statistical and the second systematic, which differs from the theoretical prediction based on isospin by 4.4σ. This indicates that there may be resonant contributions or some unknown dynamics in this decay.
We report results on an elastic cross section measurement in proton-proton collisions at a center-of-mass energy s√=510 GeV, obtained with the Roman Pot setup of the STAR experiment at the Relativistic Heavy Ion Collider (RHIC). The elastic differential cross section is measured in the four-momentum transfer squared range 0.23≤−t≤0.67 GeV2. We find that a constant slope B does not fit the data in the aforementioned t range, and we obtain a much better fit using a second-order polynomial for B(t). The t dependence of B is determined using six subintervals of t in the STAR measured t range, and is in good agreement with the phenomenological models. The measured elastic differential cross section dσ/dt agrees well with the results obtained at s√=546 GeV for proton--antiproton collisions by the UA4 experiment. We also determine that the integrated elastic cross section within the STAR t-range is σfidel=462.1±0.9(stat.)±1.1(syst.)±11.6(scale) μb.
Vor dem Hintergrund der epochalen Verunsicherungen des ausgehenden 18. Jahrhunderts stellen sich Goethe und Schiller in ihrer Dramatik den politischen Dimensionen und Abgründen der klassischen französischen Tragödie. Verkappte Staatsgründungen beherrschen ebenso das Geschehen wie der Staatsstreich, der Bürgerkrieg oder der failing state. Eine neue Aktualität gewann das Drama Corneilles und Racines jedoch erst wegen seiner Aufladung der klassischen Form mit staatspolitischen Fundamentalfragen. Das Ringen um eine Regelpoetik und ihre angemessene Umsetzung, vermeintliche Nebensächlichkeiten wie die Einheit der Zeit werden zum Schauplatz genuin politischer Reflexion. Goethe und Schiller verzichten auf eine Restauration der klassischen Form, sie entdecken aber ihre ursprüngliche Energie wieder und verhandeln diese neu. Besonders deutlich zeigt sich dies am dramaturgischen Konflikt zwischen König und Held, der einen bislang weithin übersehenen Grundpfeiler der klassischen Tragödie bildet.
The ALICE experiment at the LHC investigates the properties of the hot and dense nuclear matter created in heavy-ion collisions. By comparing the particle production in pp and p-Pb collisions, possible nuclear initial state effects can be isolated. Measurements of the ω meson pT-spectra in pp and p-Pb collisions not only allow for a determination of the nuclear modification factor RpPb, but also provide insight into the fragmentation process and serve as vital input for decay background simulations for direct photons. In this contribution, measurements of the ω meson production in pp and p-Pb collisions at √sNN=5.02 TeV are presented. This includes the signal extraction and various corrections of the ω meson yields, leading to their production cross sections and the first measured nuclear modification factor RpPb of the ω meson at LHC energies.

• Solidification and cooling of an intruded dyke or sill within the middle or shallow crust generate stresses of order 200 MPa, which relax on time scales of a few years to million years.

Abstract
Rapid emplacement of a mafic dyke or sill at mid-crustal depth heats and possibly melts the felsic wall rock followed by solidification. Associated volume changes generate stresses, possibly enforcing brittle failure and melt migration. We model the evolution of melting, solidification, temperature, and stress including visco-elastic relaxation in 1D - dykes or -sills using realistic rock rheologies of the Weschnitz pluton (Odenwald). For deep emplacement (Case 1, 15.3 km) extensive contact melting of the wall rock occurs, for shallow emplacement (Case 2, 10 km) it is negligible. The stresses are zero at high melt fractions, but increase during solidification and cooling: The intrusion orthogonal stress is always zero. The intrusion parallel stress σ‖ within the intrusion is tensile (O(200 MPa)). It relaxes on a time scale between a few years (Case 1) and 0.6 m.y. (Case 2). Within the wall rock σ‖ is compressive during heating, but becomes tensile under solidification and cooling. Wall rock stresses relax on a time scale of months to 100 years. A Deborah number is defined based on viscous to thermal relaxation allowing generalization of our results. Adding lithostatic stresses, the total stresses of Case 1 remain below the brittle strength, while for Case 2 they may exceed it. Adding the lithostatic pressure to the melt pressure, the effective stresses exceed the brittle strength and intrusion orthogonal tensile fractures are predicted. Combined with the pressure gradient within the over-pressurized felsic melts generated in the wall rock, this explains the migration of felsic contact melt into shrinkage cracks of the mafic sill in the Weschnitz pluton.
Der Beitrag thematisiert die literarische Darstellung virtueller Investigationen, welche eine performativ-schauspielerische Wiederholung von Tatgeschehnissen beinhalten. Er analysiert insbesondere die Rolle von Zufallsmotiven und -aspekten in solchen Darstellungen und deren metapoetische Bedeutung im Rahmen einer Gattungspoetik des Kriminalromans. Als Fallbeispiele dienen hierfür Friedrich Dürrenmatts 'Requiem auf den Kriminalroman' "Das Versprechen" und Stanisław Lems Roman "Der Schnupfen". Im Vergleich der beiden Texte zeigt sich unter anderem, dass die Idee eines genauen 're-enactment' von Tathergängen auf der Grundlage virtueller Rekonstruktionen in beiden Romanplots nicht direkt zum gewünschten Ergebnis führt, dass aber in Lems Roman eine Integration des Faktors Zufall in virtuelle Ermittlungsprozesse und in die Poetik des Kriminalromans insgesamt sehr viel positiver gewertet wird, während der Einfluss des Zufalls bei Dürrenmatt ebenfalls als unhintergehbar, aber nicht als uneingeschränkt begrüßenswert perspektiviert wird.
Natural Language Processing (NLP) for big data requires an efficient and sophisticated infrastructure to complete tasks both fast and correctly. Providing an intuitive and lightweight interaction with a framework that abstracts and simplifies complex tasks assists in reaching this goal. This bachelor thesis extends the NLP framework Docker Unified UIMA Interface (DUUI) by an API and a web-based graphical user interface to control and manage pipelines for automated analysis of large quantities of natural language. The extension aims to reduce the entry barrier into the field as well as to accelerate the creation and management of pipelines according to UIMA standards. Pipelines can be executed in the browser or using the web API directly and then monitored on a document level. The evaluation in usability and user experience indicates that the implementation benefits the framework by making its usage more user friendly, lightweight, and intuitive while also making the management of pipelines more efficient.
The differential cross section for 𝑍0 production, measured as a function of the boson’s transverse momentum (𝑝T), provides important constraints on the evolution of the transverse momentum dependent parton distribution functions (TMDs). The transverse single spin asymmetry (TSSA) of the 𝑍0 is sensitive to one of the polarized TMDs, the Sivers function, which is predicted to have the opposite sign in 𝑝 + 𝑝 → 𝑊 ∕𝑍 + 𝑋 from that which enters in semi-inclusive deep inelastic scattering. In this Letter, the STAR Collaboration reports the first measurement of the 𝑍0∕𝛾∗ differential cross section as a function of its 𝑝T in 𝑝+𝑝 collisions at a center-of-mass energy of 510 GeV, together with the 𝑍0∕𝛾∗ total cross section. We also report the measurement of 𝑍0∕𝛾∗ TSSA in transversely polarized 𝑝+𝑝 collisions at 510 GeV.
The dynamics of the torsion field is analyzed in the framework of the Covariant Canonical Gauge Theory of Gravity (CCGG), a De Donder–Weyl Hamiltonian formulation of gauge gravity. The action is quadratic in both, the torsion and the Riemann–Cartan tensor. Since the latter adds the derivative of torsion to the equations of motion, torsion is no longer identical to spin density, as in the Einstein–Cartan theory, but an additional propagating degree of freedom. As torsion turns out to be totally anti-symmetric, it can be parametrised via a single axial vector. It is shown in this paper that, in the weak torsion limit, the axial vector obeys a wave equation with an effective mass term which is partially dependent on the scalar curvature. The source of torsion is thereby given by the fermion axial current which is the net fermionic spin density of the system. Possible measurable effects and approaches to experimental analysis are addressed. For example, neutron star mergers could act as a dipoles or quadrupoles for torsional radiation, and an analysis of radiation of pulsars could lead to a detection of torsion wave background radiation.
Autophagy is an important degradation pathway mediating the engulfment of cellular material (cargo) into autophagosomes followed by degradation in autophagosomes.
Different stress stimuli, e.g. nutrient deprivation, oxidative stress or organelle damage, engage autophagy to maintain cellular homeostasis, recycle nutrients or remove damaged cell organelles. Autophagy not only degrades bulk cytoplasmic material but also selective autophagic cargo, for example lysosomes (lysophagy), mitochondria (mitophagy), ER (ER-phagy), lipid droplets (lipophagy), protein aggregates (aggrephagy) or pathogens (xenophagy). Selective autophagy pathways are regulated by selective autophagy receptors which bind to ubiquitinated cargo proteins and link them to LC3 on the autophagosomal membrane.
Ubiquitination is an essential post-translational modification controlling different cellular processes such as proteasomal and lysosomal degradation or innate immune signaling.
M1-linked (linear) poly-Ubiquitin (poly-Ub) chains are exclusively assembled by the E3 ligase linear ubiquitin chain assembly complex (LUBAC) and removed by the M1 poly-Ub-specific OTU domain-containing deubiquitinase with linear linkage specificity (OTULIN). In addition to key functions in innate immune signaling and nuclear factor-κB (NF-κB) activation, M1 ubiquitination is also implicated in the regulation of autophagy.
LUBAC and OTULIN control autophagy initiation and maturation and the autophagic clearance of invading bacteria via xenophagy. However, additional functions of LUBAC- and OTULIN-regulated M1 ubiquitination in autophagy are largely unknown and it also remains unexplored if LUBAC and OTULIN control other selective autophagy pathways in addition to xenophagy. This study aimed to unravel the role of LUBAC- and OTULIN-controlled M1 ubiquitination in bulk and selective autophagy in more detail.
In this study, characterization of OTULIN-depleted MZ-54 glioblastoma (GBM) cells revealed that OTULIN deficiency results in enhanced LC3 lipidation in response to autophagy induction and upon blockade of late stage autophagy with Bafilomycin A1 (BafA1). Furthermore, electron microscopy analysis showed that OTULIN-deficient cells have an increased number of degradative compartments (DGCs), confirming enhanced autophagy activity upon loss of OTULIN. APEX2-based autophagosome content profiling identified various OTULIN-dependent autophagy cargo proteins. Among these were the autophagy receptor TAX1BP1 which regulates different forms of selective autophagy (e.g. lysophagy, aggrephagy) and the glycan-binding protein galectin-3 which serves key functions in lysophagy, suggesting a role of OTULIN and M1 poly-Ub in the regulation of aggrephagy and lysophagy.

To study aggrephagy, protein aggregation was induced with puromycin which causes premature termination of translation and accumulation of defective ribosomal products (DRiPs). Loss of OTULIN increased the number of M1 poly-Ub-positive foci and insoluble proteins and reduced the levels of soluble TAX1BP1 and p62 in response to puromycin-induced proteotoxic stress.
Intriguingly, upon induction of lysosomal membrane permeabilization (LMP) with the lysosomotropic drug L-Leucyl-L-Leucine methyl ester (LLOMe), M1 poly-Ub strongly accumulated at damaged lysosomes and colocalized with TAX1BP1- and galectin-3-positive puncta. M1 poly-Ub-modified lysosomes formed a platform for NF-κB essential modulator (NEMO) and inhibitor of κB (IκB) kinase (IKK) complex recruitment and local NF-κB activation in a K63 poly-Ub- and OTULIN-dependent manner. Furthermore, inhibition of lysosomal degradation enhanced LLOMe-induced cell death, suggesting pro-survival functions of lysophagy following LMP. Enrichment of M1 poly-Ub at damaged lysosomes was also observed in human dopaminergic neurons and in primary mouse embryonic cortical neurons, confirming the importance of M1 poly-Ub in the response to lysosomal damage.
Together, these results identify OTULIN as a negative regulator of autophagy induction and the autophagic flux and reveal OTULIN-dependent autophagy cargo proteins.
Furthermore, this study uncovers novel and important roles of M1 poly-Ub in the response to lysosomal damage and local NF-κB activation at damaged lysosomes.

Abstract
We study the impact of immigrant legalization on fiscal transfers from state to local governments in the United States, exploiting variation in legal status from the 1986 Immigration Reform and Control Act (IRCA). State governments allocate more resources to IRCA counties, an allocation that is responsive to the electoral incentives of the governor. Importantly, the effect emerges prior to the enfranchisement of the IRCA migrants and we argue it is driven by the IRCA’s capacity to politically empower already legal Hispanic migrants in mixed legal status communities. The IRCA increases turnout in large Hispanic communities as well as Hispanic political engagement, without detectably triggering anti-migrant sentiment.
Although, during the past decades, substantial advances emerged in identifying major local and systemic factors contributing to initiation and progression of osteoarthritis (OA), some neuroendocrine mechanisms are still not understood or even neglected when thinking about novel therapeutic options. One of which is the sympathetic nervous system that exhibits various OA-promoting effects in different tissues of the joint. Interestingly, the β2-adrenoceptor (AR) mediates the majority of these effects as demonstrated by several in vitro, in vivo as well as in clinical studies. This review article does not only summarize studies of the past two decades demonstrating that the β2-AR plays an OA-promoting role in different tissues of the joint but also aims to encourage the reader to think about next-level research to discover novel and innovative preventive and/or therapeutic strategies targeting the β2-AR in OA.
We report a new measurement of transverse single-spin asymmetries for dijet production in collisions of polarized protons at s√ = 200 GeV. Correlations between the proton spin and the transverse momenta of its partons, each perpendicular to the proton momentum direction, are probed at high Q2 ≈160 GeV2. The associated Sivers observable ⟨kT⟩, the average parton transverse momentum, is extracted using simple kinematics. Nonzero Sivers effects are observed for the first time in dijets from proton-proton collisions, but only when the jets are sorted by their net charge, which enhances the u- or d-quark contributions to separate data samples. This also enables a simple kinematic approach for determination of the individual partonic contributions to the observed asymmetries.
We report the first measurements of cumulants, up to 4𝑡ℎ order, of deuteron number distributions and protondeuteron correlations in Au+Au collisions recorded by the STAR experiment in phase-I of Beam Energy Scan (BES) program at the Relativistic Heavy Ion Collider. Deuteron cumulants, their ratios, and proton-deuteron mixed cumulants are presented for different collision centralities covering a range of center-of-mass energy per nucleon pair √𝑠NN = 7.7 to 200 GeV. It is found that the cumulant ratios at lower collision energies favor a canonical ensemble over a grand canonical ensemble in thermal models. An anti-correlation between proton and deuteron multiplicity is observed across all collision energies and centralities, consistent with the expectation from global baryon number conservation. The UrQMD model coupled with a phase-space coalescence mechanism qualitatively reproduces the collision-energy dependence of cumulant ratios and proton-deuteron correlations.

Dynamic imaging of landmark organelles, such as nuclei, cell membrane, nuclear envelope, and lipid droplets enables image-based phenotyping of functional states of cells. Multispectral fluorescent imaging of landmark organelles requires labor-intensive labeling, limits throughput, and compromises cell health. Virtual staining of label-free images with deep neural networks is an emerging solution for this problem. Multiplexed imaging of cellular landmarks from scattered light and subsequent demultiplexing with virtual staining saves the light spectrum for imaging additional molecular reporters, photomanipulation, or other tasks. Published approaches for virtual staining of landmark organelles are fragile in the presence of nuisance variations in imaging, culture conditions, and cell types. This paper reports model training protocols for virtual staining of nuclei and membranes robust to label-free imaging parameters, cell states, and cell types. We developed a flexible and scalable convolutional architecture, named UNeXt2, for supervised training and self-supervised pre-training. The strategies we report here enable robust virtual staining of nuclei and cell membranes in multiple cell types, including neuromasts of zebrafish, across a range of imaging conditions. We assess the models by comparing the intensity, segmentations, and application-specific measurements obtained from virtually stained and experimentally stained nuclei and membranes. The models rescue the missing label, non-uniform expression of labels, and photobleaching. We share three pre-trained models, named VSCyto3D, VSCyto2D, and VSNeuromast, as well as VisCy, a PyTorch-based pipeline for training, inference, and deployment that leverages the modern OME-Zarr format.
Cyclin CLB2 mRNA localization and protein synthesis link cell cycle progression to bud growth

Clb2 is a conserved mitotic B-type cyclin, the levels of which are finely controlled to drive progression through the cell cycle. While it is known that CLB2 transcription and Clb2 protein degradation are important for precise control of its expression, it remains unclear whether the synthesis of Clb2 is also regulated. To address whether and how Clb2 expression levels respond to cell growth changes and adapt cell cycle progression, we combined single-cell and single-molecule imaging methods to measure CLB2 mRNA and protein expression throughout the Saccharomyces cerevisiae cell cycle. We found that the CLB2 mRNA was efficiently localized to the yeast bud as soon as this compartment was formed, but strikingly the Clb2 protein accumulated in the mother nucleus. The CLB2 mRNA localization in the yeast bud by the She2-3 complex did not control protein localization but rather promoted CLB2 translation. Moreover, CLB2 mRNA bud localization and protein synthesis were coupled and dependent on a single secondary structure -a ZIP code-located in the coding sequence. In a CLB2 ZIP code mutant, mRNA localization was impaired and Clb2 protein synthesis decreased, resulting in changes in cell cycle distribution and increased size of daughter cells at birth. Finally, while in WT cells the Clb2 protein concentration followed bud growth, this relationship was impaired in the ZIP code mutant. We propose that S. cerevisiae couples the control of CLB2 mRNA bud localization and protein synthesis to coordinate cell growth and cell cycle progression. This mechanism extends our knowledge of CLB2 expression regulation, and constitutes a novel function for mRNA localization.
Im Rahmen dieser Doktorarbeit werden drei Schwerpunkte behandelt: 1) Die hocheffektive Beschleunigung von Elektronen und Protonen durch die Wechselwirkung von relativistischen Laserpulsen mit Schäumen. 2) Die Erzeugung und Messung hochintensiver Betatronstrahlung von direkt laserbeschleunigten (DLA-) Elektronen. 3) Die Anwendung von DLA-Elektronen für den biologischen FLASH-Effekt mit einer rekordbrechenden Dosisrate.
Die direkte Laserbeschleunigung von Elektronen wurde durch die Wechselwirkung eines sub-ps-Laserpulses mit einer Intensität von ~ 10^19 W/cm^2 mit einem Plasma nahe kritischer Elektronendichte (NCD) untersucht. Ein sub-mm langes NCD-Plasma wurde durch Erhitzen eines Schaums mit einer niedrigen Dichte mit einem ns-Puls von 10^13-10^14 W/cm^2 erzeugt. Die Experimente wurden an der PHELIX-Anlage (Petawatt Hoch- Energie Laser für Schwerionenexperimente) in den Jahren 2019 – 2023 durchgeführt. Während der Suche nach optimalen Bedingungen für die Beschleunigung von Elektronen und Protonen wurden die Parameter des ns-Pulses variiert und verschiedene Targets verwendet. Es wurde gezeigt, dass das Plasma im Schaum gute Voraussetzungen für die Erzeugung gerichteter, ultrarelativistischer DLA-Elektronen mit Energien von bis zu 100 MeV bietet. Die Elektronen weisen eine Boltzmann-ähnliche Energieverteilung mit einer Temperatur von 10-20 MeV auf.
Optimale Bedingungen für eine effektive Beschleunigung von DLA-Elektronen wurden bei der Kombination eines CHO-Schaums mit einer Dichte von 2 mg/cm3 und einer Dicke von 300-500 µm mit einer Metallfolie erreicht. Die Gesamtladung der detektierten Elektronen mit Energien über 1,5 MeV erreichte 0,5-1 µC mit der Umwandlungseffizienz der Laserenergie von ~ 20-30%.
Außerdem wird die Beschleunigung von Protonen durch DLA-Elektronen anders verursacht als bei typischer Target Normal Sheath Acceleration (TNSA). Für die Untersuchung der lokalen Protonenenergieverteilung wurden Magnetspektrometer unter verschiedenen Winkeln zur Laserachse verwendet. Dafür wurde eine Filtermethode entwickelt, welche es ermöglicht, Spektren von Protonen mit Energien von bis zu 100 MeV zu rekonstruieren. Es wurde gezeigt, dass am PHELIX durch die Kombination von einem ~ 300-400 µm dicken CHO-Schaum mit einer Dichte von 2 mg/cm^3 und einer 10 µm dicken Au-Folie bei einer Intensität des sub-ps-Pulses von ~ 10^19 W/cm^2 und unter Verwendung eines optimierten ns-Vorpulses eine optimale Protonenbeschleunigung erreicht wurde. Es wurde ein TNSA-ähnliches Regime mit einer maximalen Cut-off-Energie von 34±0,5 MeV beobachtet. Im Vergleich dazu wurde bei der typischen TNSA unter Verwendung einer 10 µm dicken Au-Folie als Target und derselben Laserintensität eine maximale Cut-off-Energie von 24±0,5 MeV gemessen. Darüber hinaus beobachteten wir einen sehr schwachen Abfall der Protonenanzahl in Abhängigkeit von der Protonenenergie (anders als bei der typischen TNSA) und eine sehr regelmäßige Protonenstrahlverteilung in einem breiten Winkelbereich bis zu hohen Energien. Dies könnte zur Verbesserung der Qualität der Protonenradiographie von Plasmafeldern genutzt werden.
Beim DLA-Prozess (im NCD-Plasma) entsteht Betatronstrahlung durch die Oszillationen von Elektronen in quasi-statischen elektrischen und magnetischen Feldern des Plasmakanals. Um diese Strahlung zu untersuchen, wurde ein neues modifiziertes Magnetspektrometer (X-MS) konstruiert. Das X-MS ermöglicht die 1D-Auflösung mehrerer Quellen. Dank dieser Spezifikation war es möglich, Betatronstrahlung von Bremsstrahlung der ponderomotorischen Elektronen im Metallhalter zu trennen und zu messen.
Im Experiment mit einem CHO-Schaum mit einer Dichte von 2 mg/cm^3 und einer Dicke von ~ 800 µm als Target wurde die von den optimierten DLA-Elektronen erzeugte Betatronstrahlung gemessen. Bei einer Peak-Intensität des dreieckigen ns-Pulses von ~ 3·10^13 W/cm^2 und des sub-ps-Pulses von ~ 10^19 W/cm^2, welcher 4±0,5 ns gegenüber dem ns-Puls verzögert war, betrug der Halbwinkel im FWHM-Bereich des Elektronenstrahls 17±2°. Unter diesen Bedingungen war die Betatronstrahlung mit einem Halbwinkel im FWHM-Bereich von 11±2° für die Photonen mit Energien über 10 keV ebenfalls gerichtet. Die Photonenanzahl mit Energien über 10 keV wurde auf etwa 3·10^10 / 3·10^11 (gerichtete Photonen / Photonen im Halbraum entlang der Laserstrahlrichtung) abgeschätzt. Die maximale Photonenanzahl pro Raumwinkel betrug ~2·10^11 photons/sr. Die Brillanz der registrierten Betatronstrahlung erreichte ~ 2·10^20 photons/s/mm^2/mrad^2/(0.1% BW) bei 10 keV.
Die Verwendung eines Hochstromstrahls aus DLA-Elektronen für die FLASH-Strahlentherapie ermöglicht das Erreichen einer Dosis von bis zu 50-70 Gy während eines sub-ps-Laserpulses. Im Jahr 2021, während der P213-Strahlzeit am PHELIX wurde der Sauerstoffkonzentrationsabfall bei der Bestrahlung von Medien (Wasser und andere biologische Medien) mit DLA-Elektronen in Abhängigkeit von der Dosis untersucht. Die Strahlendosis wurde hierbei indirekt gemessen. Hierfür wurde eine Rekonstruktionsmethode entwickelt, die es ermöglicht, die Dosis innerhalb des „Wasser-Containers“ auf Basis von Messungen außerhalb des Containers mit einem untersuchten Medium zu ermitteln. Es wurde eine gute Übereinstimmung zwischen dem Experiment und einer Monte-Carlo-Simulation für Wasser gezeigt. Die registrierte Dosisrate erreichte einen Rekordwert von ~ 70 TGy/s.
Carbonate archives record a brief snapshot of the ambient Earth’s surface conditions at their deposition. However, the geologically reasonable extraction and interpretation of geochemical proxy data from ancient, diagenetically altered rock archives is fraught with problems. Three issues stand out: the dichotomy between petrographic and geochemical alteration; the lack of quantitative age constraints for specific diagenetic phases resulting in a poorly constrained admixture of local, basin-wide and over-regional (far-field) features; and an often insufficient understanding of the temperatures and compositions of diagenetic fluids. Here, the archive of Devonian marine limestones exposed to multiple far-field diagenetic events is used as an example to explore the above-listed issues. Methods applied include petrography, micro XRF, fluid inclusion data, clumped isotopes, δ13C and δ18O isotopes, 87Sr/86Sr ratios and quartz trace element data. Devonian limestones studied here were overprinted by two cross-cutting regional fault zones (T ≈ 230 °C) by multiple events between the Variscan Orogeny and the late Paleogene. The following processes are recorded: (i) protolith deposition and partial dolomitisation during rapid burial in the Middle/Late Devonian (T ≈ 180 °C); (ii) deep burial to ca 6.5 km and tectonic/hydrothermal overprint during the Variscan Orogeny in the Carboniferous (T ≈ 90–230 °C); (iii) rapid uplift to 1–2 km burial depth at the end of the Variscan Orogeny and hypogene karstification (T ≈ 50 to 100 °C) initiated by regional geology in the Permian/Triassic; (iv) tectonic/hydrothermal overprint during the opening of the Proto-Atlantic Ocean between the Early Jurassic and the Early Cretaceous (T ≈ 50 to 130 °C); (v) tectonic/hydrothermal overprint including renewed hypogene karstification and hydrothermal calcite cement precipitation (T ≈ 50 to 180 °C) during Alpine Orogeny between the Late Cretaceous and late Paleogene. Despite this complex series of diagenetic events, the protolith limestones largely preserved their respective Middle/Late Devonian dissolved inorganic carbon (DIC) and 87Sr/86Sr signatures. This study documents that geochemical proxy data, placed into their petrographic, paleotemperature, and local to over-regional context, significantly increases the ability to extract quantitative information from ancient carbonate rock archives. Research shown here has wider relevance for carbonate archive research in general.
Das Schilddrüsenkarzinom (SK) ist die häufigste bösartige endokrine Tumorerkrankung. Während das nicht-metastasierte und nicht-mutierte papilläre Schilddrüsenkarzinom (PSK) und das follikuläre Schilddrüsenkarzinom (FSK) eine gute Heilungschance aufweisen, zeigen die mutierten und metastasierten Varianten des PSK und FSK sowie das anaplastische Schilddrüsenkarzinom (ASK) weiterhin eine schlechte Prognose. Die Entwicklung von Therapieresistenzen stellen hierbei ein Hauptproblem in der Behandlung des fortgeschrittenen Schilddrüsenkarzinoms dar.
In den letzten Jahren wurden in Studien zunehmend Tumor-initiierende Zellen (TIZ) beschrieben, welche eine kleine Subpopulation von Zellen mit der Fähigkeit zur Selbsterneuerung, Tumorinitiierung und Entwicklung von Therapieresistenzen von Tumoren darstellen. Die Existenz von TIZ wurde auch im SK nachgewiesen. Ein entscheidender Faktor für die Persistenz von TIZ ist die Hypoxie, welche über eine Veränderung des Tumormikromilieus und des Zellmetabolismus zur Entstehung von Therapieresistenzen beiträgt. Ein durch Hypoxie hochreguliertes Enzym ist die Carboanhydrase IX (CAIX). CAIX wird hauptsächlich von Tumorzellen exprimiert und katalysiert die Reaktion von Kohlendioxid zu Bicarbonat und einem Proton und trägt damit zur Säurepufferung der Tumorzelle bei. CAIX stellt somit einen entscheidenden Faktor für das Überleben von Tumorzellen in einem hypoxischen Milieu dar. Des Weiteren ist eine erhöhte Expression von CAIX mit einem schlechten Patienten-Outcome assoziiert, wie z.B. im Brustkrebs. Diese Eigenschaften machen CAIX zu einem attraktiven Angriffspunkt einer zielgerichteten Tumortherapie. Die vorliegende Studie hat zum Ziel, die Expression von CAIX sowie dessen biologische Rolle im Schilddrüsenkarzinom näher zu untersuchen.
Hierzu wurden Proben von 114 SK-Patienten immunhistochemisch auf eine CAIX-Expression untersucht und mit tumorfreiem Schilddrüsengewebe verglichen. Hierbei waren unterschiedliche SK-Subtypen vertreten. Für eine weitere Validierung der Expressionsdaten erfolgte die Auswertung eines Datasets von „The Cancer Genome Atlas“ (TCGA) mithilfe von cBioportal. Da die Hypoxie ein wichtiger Faktor für die Persistenz von TIZ ist, wurde die CAIX-Expression in Tumorsphären, ein in vitro Nachweis von TIZ-Aktivität, mittels der Durchflusszytometrie bestimmt und mit der CAIX-Expression von Monolayern verglichen. Als SK-Zelllinien wurden BCPAP (PSK), FTC 133 (FSK) und 8505 C (ASK) verwendet. Anschließend wurde mithilfe der Polymerasekettenreaktion und Immunofluoreszenzfärbung untersucht, ob eine CAIX-Expression in den Tumorsphären mit der Expression von bereits bekannten Stammzellmarkern, u.a. NANOG, assoziiert ist. Die Unterschiede der CAIX-Expression, nach Inkubation der Monolayer jeweils in Normoxie und Hypoxie, wurden mittels Durchflusszytometrie bestimmt. Mithilfe eines genetischen CAIX-Knockdowns sowie einer pharmakologischen Inhibition mit dem CAIX-Inhibitor Methazolamid (MZM) wurde die Tumorzellproliferation und -Sphärenbildung unter Normoxie und Hypoxie bestimmt. Zusätzlich wurde der Einfluss von MZM auf die Apoptose und den Zellzyklus untersucht.
Immunhistochemische Färbungen der Gewebeproben von SK-Patienten zeigten, dass die CAIX-Expression sowohl im PSK und FSK als auch im ASK im Vergleich zum tumorfreien Schilddrüsengewebe erhöht war. Des Weiteren zeigte die klinisch-pathologische Datenanalyse, dass eine erhöhte CAIX-Expression mit dem Auftreten von Lymphknotenmetastasen im differenzierten SK assoziiert war. Auch die Analyse des TCGA-Datasets bestätigte, dass eine erhöhte Expression der CAIX-mRNA mit einem fortgeschrittenen Tumorstadium, Fernmetastasen und mit einem kürzeren Gesamt-Überleben von SK-Patienten korrelierte. Die weiteren funktionellen in vitro Untersuchungen ergaben, dass die CAIX-Expression in den Tumorsphären im Vergleich zu Monolayern erhöht und mit einer erhöhten Expression von Stammzellmarkern assoziiert war. Ein genetischer CAIX-Knockdown und eine CAIX-Inhibition mit MZM führten über eine Induktion der Apoptose und eines Zellzyklusarrests zu einer verminderten Tumorzellproliferation und Sphärenbildung.
Zusammenfassend deuten die Ergebnisse darauf hin, dass CAIX ein vielversprechendes Zielmolekül für eine gezielte Tumortherapie des fortgeschrittenen SK ist. Um diese Hypothese bestätigen zu können, sind jedoch weitere prospektive Analysen von Patientenproben sowie funktionelle in vivo Untersuchungen am SK nötig.

Abstract
We conducted scaled analogue modelling to show the influence of varying single layer initial orientation on the geometry of folds and boudins in a bulk constrictional strain field. The initial angle between the plane of shortening and the competent layer (θZ(i)) was incrementally increased from 0° to 90° by multiples of 11.25°. While the amount of layer thickening decreased with increasing θZ(i), the deformation structures produced range from pure dome-and-basin folds to coeval folds and boudins. Based on the attitude of fold axes, there are extension-parallel (FEPR) and extension-perpendicular (FEPP) folds, with axes subparallel and subperpendicular to the principal stretching axis (X), respectively. Coeval growth of FEPR folds and boudins occurred when θZ(i) > ca. 25°. The FEPP folds can be subdivided into a first type which affect the entire layer (if θZ(i) ranges between 11.25 and 78.75°) and a second type, referred to as FBEPP folds, which are affecting pre-existing boudins if θZ(i) > 45°. The interlimb angle of all types of folds increases with increasing θZ(i). Folds and boudins similar to the ones produced in this study can be found in salt domes and in tectonites of subduction zones.
Rationale: Potassium (K) is a major component of several silicate minerals and seawater, and, therefore, constraining past changes in the potassium cycle is a promising way of tracing large-scale geological processes on Earth. However, [K] measurement using inductively coupled plasma mass spectrometry (ICP-MS) is challenging due to an ArH+ interference, which may be of a similar magnitude to the K+ ion beam in samples with <0.1% m/m [K].
Methods: In this work, we investigated the effect of the ArH+ interference on K/Ca data quality by comparing results from laser-ablation (LA)-ICP-MS measured in medium and high mass resolution modes and validating our LA results via solution ICP-optical emission spectroscopy (OES) and solution ICP-MS measurements. To do so, we used a wide range of geological reference materials, with a particular focus on marine carbonates, which are potential archives of past changes in the K cycle but are typically characterised by [K] < 200 μg/g. In addition, we examine the degree to which trace-element data quality is driven by downhole fractionation during LA-ICP-MS measurements.
Results: Our results show that medium mass resolution (MR) mode is sufficiently capable of minimising the effect of the ArH+ interference on K+. However, the rate of downhole fractionation for Na and K varies between different samples as a result of their differing bulk composition, resulting in matrix-specific inaccuracy. We show how this can be accounted for via downhole fractionation corrections, resulting in an accuracy of better than 1% and a long-term reproducibility (intermediate precision) of <6% (relative standard deviation) in JCp-1NP using LA-ICP-MS in MR mode.
Conclusion: Our [K] measurement protocol is demonstrably precise and accurate and applicable to a wide range of materials. The measurement of K/Ca in relatively low-[K] marine carbonates is presented here as a key example of a new application opened up by these advances.

Williams' Roman "Clifford's Blues" (1999), der in der Form eines fiktiven, von einem afroamerikanischen Jazz-Musiker geschriebenen Tagebuchs von der Inhaftierung im Konzentrationslager Dachau erzählt. Der Roman lässt sich nicht nur als mit den Mitteln der Fiktion arbeitender Beitrag zur Geschichte der Verfolgung Schwarzer Menschen im Nationalsozialismus verstehen, sondern auch als selbst transatlantisches, vermeintlich historisches Zeitzeugnis, das aus einer ungewöhnlichen Perspektive die Thematik eines unsteten Archivs verflochtener Gewalt- und Exilerfahrungen beleuchtet. Das Augenmerk der Analyse liegt einerseits auf den Chancen eines solchen Archivs: Der Roman zeigt Möglichkeiten diasporischer Gemeinschaftsbildung und beschreibt die transatlantische Perspektive des Protagonisten als Ressource für das Erkennen von Kontinuitäten der Unterdrückung wie auch für das Ableiten von alltäglichen Widerstandsstrategien. Andererseits werden ausgehend vom Kunstgriff der Herausgeberfiktion und von Dokumenten aus dem Nachlass Risiken und Widersprüche des hier entworfenen Archivs diskutiert. Es wird gezeigt, dass dieses zwar auf Transnationalität zielt, jedoch teilweise der Rückbindung an ein lokales Moment bedarf.
Non-matrix-matched calibration of laser ablation ICPMS (trace/major) element data is a common quantification strategy. However, LA sampling is associated with downhole elemental fractionation, potentially causing inaccuracies if the magnitude of fractionation between the sample and reference material (RM) differs. Here, we estimate fractionation factors (FFs) for different elements (El) in a range of RMs relative to NIST SRM610/612 (FFEl/Ca-NIST) and evaluate element-specific corrections for downhole fractionation using these measured FFEl/Ca-NIST. Significantly different mean El/Ca values were observed before and after correction, particularly for the alkali elements (all RMs), and B, Fe, and Zn (some RMs), notably improving accuracy, especially for the alkali elements. In cases where this methodology does not result in an accuracy improvement, this may help identify underlying issues in reported/reference values for RMs, given that this phenomenon should be accounted for. Overall, we recommend considering routine assessment of FFs and applying a FF correction to enhance data quality.
Der Natrium-abhängige Kaliumkanal Slack (KNa1.1, Slo2.2, KCNT1) nimmt eine Schlüsselrolle in der Regulation neuronaler Erregbarkeit ein, indem er die Ausbildung und Feuerungsfrequenz von Aktionspotentialen kontrolliert. Sowohl in Mäusen als auch in Menschen wird Slack besonders hoch in nicht-peptidergen C-Faser-Neuronen exprimiert. Wissenschaftliche Erkenntnisse der letzten Jahre konnten die Beteiligung von Slack-Kanälen in der Signalverarbeitung neuropathischer Schmerzen, aber auch in verschiedenen Arten von Pruritus, feststellen. Dabei zeigen Slack-defiziente Mäuse ein verstärktes mechanisches Schmerzverhalten nach einer peripheren Nervenverletzung und ein erhöhtes Kratzverhalten in akuten Juckreiz-Modellen. Das als Slack-Aktivator identifizierte trizyklische Neuroleptikum Loxapin zeigt sowohl analgetische als auch antipruritische Effekte in Mäusen, jedoch ist sein klinischer Einsatz auf Grund schwerwiegender antipsychotischer Nebenwirkungen limitiert. Basierend auf Loxapins Leitstruktur wurden daher in dieser Arbeit neue Slack-Aktivatoren mit einem verbesserten pharmakologischen Profil designed und ihr Potential für die Therapie von Schmerzen sowie akutem und chronischem Pruritus in vivo untersucht.
The hepatitis B virus is one of the most common causes of virus-related chronic liver disease and remains a major global health problem affecting 296 million people worldwide. Despite an available and highly effective vaccination, hepatitis B infections lead to an annual mortality rate of approximately 0.8 million people. The global prevalence is heterogeneously distributed and reflects a high infections and chronicity, particularly in low-income countries, due to a lack of vaccination strategies, underdiagnosis and low treatment rates. A complete cure remains undiscovered to this day. Based on their genetic makeup, the virus is categorized into nine genotypes with a genetic difference of more than 8% within the sequence. In addition to their geographical distribution, hepatitis B virus genotypes also differ in terms of their clinical outcome, pathogenesis and treatment response.
The viral protein HBx is known to interact with several cellular signaling pathways and is thereby accounted as the driving force in the development of hepatitis B virus-associated pathogenesis and progression of hepatocellular carcinoma. In particular, HBx interacts with mitochondria and induces profound alterations in the mitochondrial morphology and function with a severe impact on the liver’s physiology and with an emerging role in liver-related disease progression.
This study aims to investigate the genotype-related impact of HBx with regard to their interaction with cellular signaling pathways. A particular focus was placed on mitochondria-dependent interactions and signaling pathways in order to broaden the understanding of the genetic diversity of the genotypes.
Differences between genotypes of HBx were examined and compared through in vitro experiments based on a cell culture-based system. Plasmid DNA encoding the HBx protein of the different genotypes was transiently transfected into Huh7 or HepG2 cells and examined for molecular and protein-biochemical effects on the host cell, usually 72 hours after transfection. This study focused on the most common genotypes A, B, C, D, E and G worldwide.
Based on initial kinome profiling analyses, it was found that HBx differs greatly within their genetic variants and suggests different effects on overall cell function and in particular on mitochondrial kinases. Furthermore, confocal laser scanning microscopy reveals profound HBx-mediated changes in the mitochondrial network structure, however with major differences among the different genotypes. In particular, HBx of genotypes A and G causes enormous fragmentation of mitochondrial structures, accompanied by emergent changes in mitochondrial function. Due to an increased interaction with the voltage-dependent anion channel 3, a significant loss of mitochondrial membrane potential was also observed, together with an increased radial oxygen stress level and an induction of central mitochondria-dependent inflammatory mediators. In contrast, the contribution of HBx-genotype B and E reveals only moderate effects in these regards. Using a pH-sensitive reporter system, HBx genotypes which previously indicated a strong distribution in the mitochondrial morphology and function, also showed an elevated mitophagy through the PINK1/Parkin-mediated pathway. This study provides direct evidence that HBx-mediated changes in host cell signaling pathways, especially in mitochondrial-associated pathways, fundamentally dependent on the different genotypes. In addition, the results also indicate an important role of HBx in the process of genotype-dependent liver pathogenesis and provide insight into the underlying cellular mechanisms and signaling pathways.

In vier Studien werden jüdische Erfahrungen der Moderne zwischen Zeigen und Beobachten, sozialer Schau und antisemitischer Bedrängnis als Modi des Theatralen zum Thema. Dabei rückt Theater als Praxis ins Zentrum, mit der Zugehörigkeit in der Moderne verhandelt, selbstbewusst behauptet oder befragt wurde. Während sich Juden und Jüdinnen "wie auf einer Bühne" permanenter Beobachtung ausgesetzt sahen, kommentierten sie dies wiederum theatral, erprobten eigene Strategien sozialer Teilhabe und beteiligten sich an der Ausgestaltung von bürgerlichen Sozialrollen in der Großstadt. Theater zeigt sich so als vielschichtige kulturelle Praxis wie als "Spiel- und Spiegelform des Lebens" (Stefan Zweig).
Stimulated emission depletion (STED) microscopy is a super-resolution technique that surpasses the diffraction limit and has contributed to the study of dynamic processes in living cells. However, high laser intensities induce fluorophore photobleaching and sample phototoxicity, limiting the number of fluorescence images obtainable from a living cell. Here, we address these challenges by using ultra-low irradiation intensities and a neural network for image restoration, enabling extensive imaging of single living cells. The endoplasmic reticulum (ER) was chosen as the target structure due to its dynamic nature over short and long timescales. The reduced irradiation intensity combined with denoising permitted continuous ER dynamics observation in living cells for up to 7 hours with a temporal resolution of seconds. This allowed for quantitative analysis of ER structural features over short (seconds) and long (hours) timescales within the same cell, and enabled fast 3D live-cell STED microscopy. Overall, the combination of ultra-low irradiation with image restoration enables comprehensive analysis of organelle dynamics over extended periods in living cells.
Correlative dynamic imaging of cellular landmarks, such as nuclei and nucleoli, cell membranes, nuclear envelope and lipid droplets is critical for systems cell biology and drug discovery, but challenging to achieve with molecular labels. Virtual staining of label-free images with deep neural networks is an emerging solution for correlative dynamic imaging. Multiplexed imaging of cellular landmarks from scattered light and subsequent demultiplexing with virtual staining leaves the light spectrum for imaging additional molecular reporters, photomanipulation, or other tasks. Current approaches for virtual staining of landmark organelles are fragile in the presence of nuisance variations in imaging, culture conditions, and cell types. We report training protocols for virtual staining of nuclei and membranes robust to variations in imaging parameters, cell states, and cell types. We describe a flexible and scalable convolutional architecture, UNeXt2, for supervised training and self-supervised pre-training. The strategies we report here enable robust virtual staining of nuclei and cell membranes in multiple cell types, including human cell lines, neuromasts of zebrafish and stem cell (iPSC)-derived neurons, across a range of imaging conditions. We assess the models by comparing the intensity, segmentations, and application-specific measurements obtained from virtually stained and experimentally stained nuclei and cell membranes. The models rescue missing labels, non-uniform expression of labels, and photobleaching. We share three pre-trained models (VSCyto3D, VSNeuromast, and VSCyto2D) and a PyTorch-based pipeline (VisCy) for training, inference, and deployment that leverages current community standards for image data and metadata.
Snapshots of acetyl-CoA synthesis, the final step of CO₂ fixation in the Wood-Ljungdahl pathway

In the ancient microbial Wood-Ljungdahl pathway, CO2 is fixed in a multi-step process with acetyl-CoA synthesis at the bifunctional carbon monoxide dehydrogenase/acetyl-CoA synthase complex (CODH/ACS). Here, we present catalytic snapshots of the CODH/ACS from the gas-converting acetogen Clostridium autoethanogenum, characterizing the molecular choreography of the overall reaction including electron transfer to the CODH for CO2 reduction, methyl transfer from the corrinoid iron-sulfur protein (CoFeSP) partner to the ACS active site and acetyl-CoA production. Unlike CODH, the multidomain ACS undergoes large conformational changes to form an internal connection to the CODH active site, accommodate the CoFeSP for methyl transfer and protect the reaction intermediates. Altogether, the structures allow us to draw a detailed reaction mechanism of this enzyme crucial for CO2 fixation in anaerobic organisms.
By analyzing (27.12±0.14)×108 ψ(3686) events accumulated with the BESIII detector, the decay ηc(2S)→K+K−η is observed for the first time with a significance of 6.2σ after considering systematic uncertainties. The product of the branching fractions of ψ(3686)→γηc(2S) and ηc(2S)→K+K−η is measured to be B(ψ(3686)→γηc(2S))×B(ηc(2S)→K+K−η)=(2.39±0.32±0.34)×10−6, where the first uncertainty is statistical, and the second one is systematic. The branching fraction of ηc(2S)→K+K−η is determined to be B(ηc(2S)→K+K−η)=(3.42±0.46±0.48±2.44)×10−3, where the third uncertainty is due to the branching fraction of ψ(3686)→γηc(2S). Using a recent BESIII measurement of B(ηc(2S)→K+K−π0), we also determine the ratio between the branching fractions of ηc(2S)→K+K−η and ηc(2S)→K+K−π0 to be 1.49±0.22±0.25, which is consistent with the previous result of BaBar at a comparable precision level.
Search for X(3872)→π⁰π⁰χc₁,₂

Using 10.1 fb−1 of e+e− collision data collected by the BESIII detector with center-of-mass energies between 4.15 GeV and 4.30 GeV, we search for the decays X(3872)→π0π0χc1,2, where the X(3872) is produced in e+e−→γX(3872). No evidence above 3σ is found for either decay. Upper limits at the 90% C.L. on the branching fractions of X(3872)→π0π0χc1,2 normalized to the branching fraction of X(3872)→π+π−J/ψ are set to be B(X(3872)→π0π0χc1)/B(X(3872)→π+π−J/ψ)<1.1 and B(X(3872)→π0π0χc2)/B(X(3872)→π+π−J/ψ)<0.5, taking into account both statistical and systematic uncertainties.
