Der Hype um WikiLeaks ist verpufft – das wurde bereits in der Begrüßung von Prof. Dr. Nicole Deitelhoff, Prof. Dr. Christopher Daase und Dr. Thorsten Thiel von der Universität Frankfurt klar. Doch eine Reihe ungeklärter Fragen und Probleme rund um das Prinzip Leaking bleiben zurück. Um diese zu diskutieren, begaben sich am vergangenen Mittwoch einige Interessierte zum Workshop “Leaking: Sicherheitsbedrohung oder subversive Demokratisierung?” an die Goethe-Universität Frankfurt. Dabei sollte es um grundlegende, wissenschaftliche Fragen gehen, die im Hype um WikiLeaks und Julian Assange oftmals untergegangen sind: Wodurch werden Leaks legitimiert und wo verläuft die Grenze zu Illegitimität? Was ist die politische Funktion des Leaking? Können oder müssen Leaks neutral sein? Und wie gefährlich sind sie für Sicherheitspolitik?...
Chromosomal translocations can lead to the formation of chimeric genes encoding fusion proteins such as PML/RARalpha, PLZF/RARalpha, and AML-1/ETO, which are able to induce and maintain acute myeloid leukemia (AML). One key mechanism in leukemogenesis is increased self renewal of leukemic stem cells via aberrant activation of the Wnt signaling pathway. Either X-RAR, PML/RARalpha and PLZF/RARalpha or AML-1/ETO activate Wnt signaling by upregulating gamma-catenin and beta-catenin. In a prospective study, a lower risk of leukemia was observed with aspirin use, which is consistent with numerous studies reporting an inverse association of aspirin with other cancers. Furthermore, a reduction in leukemia risk was associated with use of non-steroidal anti-inflammatory drug (NSAID), where the effects on AML risk was FAB subtype-specific. To better investigate whether NSAID treatment is effective, we used Sulindac Sulfide in X-RARalpha-positive progenitor cell models. Sulindac Sulfide (SSi) is a derivative of Sulindac, a NSAID known to inactivate Wnt signaling. We found that SSi downregulated both beta-catenin and gamma-catenin in X-RARalpha-expressing cells and reversed the leukemic phenotype by reducing stem cell capacity and increasing differentiation potential in X-RARalpha-positive HSCs. The data presented herein show that SSi inhibits the leukemic cell growth as well as hematopoietic progenitors cells (HPCs) expressing PML/RARalpha, and it indicates that Sulindac is a valid molecular therapeutic approach that should be further validated using in vivo leukemia models and in clinical settings.
Neolecanium amazonensis Foldi is redescribed and illustrated and is transferred to the new genus Foldilecanium Kondo as Foldilecanium amazonensis (Foldi) comb. nov. A new species, Foldilecanium multisetosus Kondo, is described and illustrated based on specimens collected in Cali, Colombia, on Cananga odorata (Lam.) Hook.f. and Thomson (Annonaceae). An updated taxonomic key to New World Myzolecaniinae and a key to separate the two species of Foldilecanium are provided.
Virus-infected cells are eliminated by cytotoxic T lymphocytes, which recognize viral epitopes displayed on major histocompatibility complex class I molecules at the cell surface. Herpesviruses have evolved sophisticated strategies to escape this immune surveillance. During the lytic phase of EBV infection, the viral factor BNLF2a interferes with antigen processing by preventing peptide loading of major histocompatibility complex class I molecules. Here we reveal details of the inhibition mechanism of this EBV protein. We demonstrate that BNLF2a acts as a tail-anchored protein, exploiting the mammalian Asna-1/WRB (Get3/Get1) machinery for posttranslational insertion into the endoplasmic reticulum membrane, where it subsequently blocks antigen translocation by the transporter associated with antigen processing (TAP). BNLF2a binds directly to the core TAP complex arresting the ATP-binding cassette transporter in a transport-incompetent conformation. The inhibition mechanism of EBV BNLF2a is distinct and mutually exclusive of other viral TAP inhibitors.
Das onkogene Fusionsprotein AML1/ETO entsteht durch die chromosomale Translokation t(8;21), die in etwa 12 % aller primären akuten myeloischen Leukämien (AML) auftritt. Die DNA-Bindedomäne des hämatopoetischen Transkriptionsfaktors AML1 wird hierbei mit fast dem gesamten ETO-Protein fusioniert, das als transkriptioneller Repressor wirkt. In den transformierten Zellen kommt es somit zur Blockierung der myeloischen Differenzierung und zur verstärkten Proliferation. Entscheidend für das leukämische Potential von AML1/ETO ist die Fähigkeit zur Oligomerisierung, die durch die Nervy-Homologie-Region-2 (NHR2)-Domäne im ETO-Anteil vermittelt wird.
Durch lentivirale Transduktion konnte bereits gezeigt werden, dass Proteine, welche die NHR2-Domäne enthalten, die Oligomerisierung von AML1/ETO inhibieren und damit den leukämischen Phänotyp AML1/ETO-exprimierender myeloischer Zellen aufheben. In der vorliegenden Arbeit sollten nun alternative Wege zur Einbringung der therapeutischen Proteine in t(8;21)-positive AML-Zellen untersucht werden. Dafür wurde sowohl die Möglichkeit der Proteintransduktion als auch die Verwendung nicht-integrierender viraler Vektoren analysiert.
Im ersten Projekt wurden durch Fusion mit der HIV-1 TAT-Domäne zellpermeable NHR2-Proteine generiert. Zunächst wurde ein Protokoll zur Expression und Reinigung der rekombinanten Proteine etabliert. Durch eine ausführliche biochemische Charakterisierung konnte gezeigt werden, dass die aus Bakterien aufgereinigten NHR2-Proteine funktionell und sehr rein waren. Sie wiesen den erwarteten hohen alpha-helikalen Anteil auf und behielten ihre Fähigkeit zur Bildung von Tetrameren in vitro bei. Die TAT-NHR2-Fusionsproteine sind in der Lage, in humane Zellen einzudringen und konnten erfolgreich in den Lysaten nachgewiesen werden. Mikroskopische Studien zeigten, dass der Großteil der internalisierten Proteine in Endosomen-ähnlichen Vesikeln lokalisiert war. Die Zugabe des Endosomeninhibitors Chloroquin oder eines endosomolytischen, zellpermeablen Peptides ermöglichte eine erhöhte intrazelluläre Stabilität der zellpenetrierenden Proteine. Co-Immunpräzipitations-Experimente konnten bestätigen, dass die aufgenommenen NHR2-Proteine spezifisch an das ETO-Protein in transfizierten, adhärenten Zellen binden können. Die Proteintransduktion in die myeloische, AML1/ETO-wachstumsabhängige Zelllinie Kasumi-1 ist unter serumfreien Bedingungen ebenfalls möglich. Die konsekutive Behandlung der AML-Zellen mit den TAT-NHR2-Fusionsproteinen führte zu einer Reduktion der Expression des Stammzellmarkers c-kit (CD117) in 26 % der behandelten Zellen. Die Anwendung zellpermeabler NHR2-Proteine ist demnach prinzipiell möglich, bedarf aber weiterer Optimierung, um die notwendige hohe Bioverfügbarkeit zu erreichen.
In einem zweiten Projekt wurden Adeno-assoziierte virale (AAV) Vektoren verwendet, um die NHR2-Proteine in den hämatopoetischen Zellen zu exprimieren. Mit Hilfe mehrerer Methoden konnte gezeigt werden, dass sich mit den generierten Vektoren, die auf dem AAV-Serotyp 2 basierten, erfolgreich eine transiente Genexpression induzieren ließ. Der CMV-Promoter vermittelte jedoch nur eine schwache Expression in den hämatopoetischen Zellen. Unter Verwendung des stärkeren SFFV-Promoters konnte die Expressionsstärke deutlich gesteigert werden. Die von den optimierten AAV-Vektoren vermittelte Expression der NHR2-Proteine führte in den beiden AML1/ETO-positiven Zelllinien Kasumi-1 und SKNO-1 zu den erwarteten, spezifischen Effekten. So wurde das Wachstum verlangsamt und gleichzeitig die Apoptoserate erhöht. AML1/ETO-unabhängige Zellen wurden dagegen von den AAV-NHR2-Vektoren nicht beeinflusst. Obwohl die Proteinexpression in SKNO-1 Zellen stärker war, zeigten die Kasumi-1 Zellen deutlichere Effekte. Die NHR2-Proteine bewirkten in den transduzierten t(8;21)-positiven Zellen außerdem eine Reduktion der Expression der Stammzellmarker CD34 bzw. c-kit. Dies deutet auf eine partielle Differenzierung der beiden AML1/ETO-abhängigen Zelllinien hin. Damit ließen sich durch AAV-vermittelte Transduktion in den AML-Zellen dieselbe Wirkung in Hinblick auf Wachstum, Differenzierbarkeit und Apoptoserate erzielen wie dies mit den lentiviralen Vektoren zuvor beschrieben wurde. In einem abschließenden Vergleich wurde aber deutlich, dass nicht-integrierende Vektorsysteme generell eine schwächere NHR2-Proteinexpression induzieren und demzufolge auch schwächere Effekte als integrierende Vektoren in den AML1/ETO-positiven Zellen auslösen.
A thick Middle and Late Pleistocene loess/palaeosol sequence is exposed at the gravel quarry Gaul located east of Weilbach in the southern foreland of the Taunus Mountains. The loess/palaeosol sequence correlates to the last three glacial cycles. Seven samples were dated by luminescence methods using an elevated temperature IRSL (post-IR IRSL) protocol for polymineral fine-grains to determine the deposition age of the sediment and to set up a more reliable chronological framework for these deposits. The fading corrected IR50 and the pIRIR225 age estimates show a good agreement for almost all samples. The fading corrected IRSL ages range from 23.7 ± 1.6 ka to >350 ka indicating that the oldest loess was deposited during marine isotope stage (MIS) 10 or earlier and that the humic-rich horizon (Weilbacher Humuszone) was developed during the late phase of MIS 7. Loess taken above the fCc horizon most likely accumulated during MIS 6 indicating that the remains of the palaeosol are not belonging to the last interglacial soil. The two uppermost samples indicate that the youngest loess accumulated during MIS 2 (Upper Würmian). Age estimates for the loess-palaeosol sequence of the gravel quarry Gaul/Weilbach could be obtained up to ~350 ka using the pIRIR225 from feldspar. Seit den 1980er Jahren fällt dem Computer sowohl in Beruf und Bildung als auch in der Freizeit eine wachsende Bedeutung zu (z.B. Rheinberg & Tramp, 2006). Häufig ist es für Computernutzer erforderlich, sich Computerkenntnisse selbstständig und ohne explizite Anleitung anzueignen (Richter, Naumann & Horz, 2010). Männer und Frauen erleben dabei den Umgang mit dem Computer unterschiedlich; diese Unterschiede können in Lern- und Leistungsdifferenzen resultieren (z.B. Baloğlu & Çevik, 2008). Zur Erklärung potenzieller Lern- und Leistungsunterschiede zwischen den Geschlechtern wurde in der Arbeit das kognitiv-motivationale Prozessmodell zugrunde gelegt. Das Modell erlaubt es, Leistungsunterschiede prozessnah über die aktuelle Motivation, das Flow-Erleben und die Strategiesystematik zu erklären. In Studie 1 (N = 18) wurde ein Beobachtungssystem zur Erfassung von optimalen und suboptimalen Explorationsstrategien am Computer entwickelt. In Studie 2 (N = 33) und 3 (N = 92) wurde im Rahmen eines quasiexperimentellen Designs getestet, ob sich weibliche und männliche Studierende der Wirtschaftswissenschaften in Strategiesystematik, Motivation und Leistung beim selbstregulierten Erlernen des Programms SPSS unterscheiden. Die Teilnehmer hatten nach einer kurzen Einführung in die Benutzeroberfläche von SPSS die Aufgabe, mehrere Statistikaufgaben mit SPSS zu lösen. Die Ergebnisse der Studien erbrachten zum Teil inkonsistente Befunde: Über die Studien hinweg zeigten sich keine statistisch signifikanten Unterschiede in der Motivation vor Beginn der Aufgabenbearbeitung. In Studie 2 ergaben sich jedoch Geschlechtsunterschiede in der aktuellen Motivation und der Strategiesystematik während des Bearbeitungsprozesses sowie in der Leistung. Frauen erreichten dabei niedrigere Werte bzw. weniger Punkte. In beiden Studien standen die aktuelle Motivation, das Flow-Erleben sowie die Strategiesystematik modellkonform in Beziehung mit der Leistung. Die Ergebnisse und die Methodik der Arbeit wurden abschließend kritisch diskutiert.
A comprehensive overview of plant, fungus and animal species of Austria revealed a total of 748 endemic and subendemic species, including, 11 harvestman and 46 spider species. Altogether two endemic harvestmen (Nemastoma bidentatum relictum, Nemastoma schuelleri) and 8 endemic spiders (Abacoproeces molestus, Collinsia (caliginosa) nemenziana, Mughiphantes severus, Mughiphantes styriacus, Pelecopsis alpica, Scotophaeus nanus, Troglohyphantes novicordis, Troglohyphantes tauriscus), beside 9 subendemic harvestman and 38 subendemic spider species have been recorded from Austria. Hot-spots of endemism in the Eastern Alps are the north-eastern (Ennstaler Alps) and southern Calcareous Alps (Karawanken, Karnische Alps) and the Central Alps (Hohe Tauern, Gurktaler Alps, Ötztaler and Stubaier Alps). Most of the endemic arachnid species occur from the nival down to the montane zone. Important habitats are rocky areas, caves and woodlands. High absolute numbers and percentages of endemics can be found within the harvestman families Cladonychiidae, Ischyropsalididae and Nemastomatidae and in the spider genera Lepthyphantes s. l. and Troglohyphantes. The conservation status of these highly endangered taxa – 85 % of the spider species and 100 % of the harvestman taxa are endangered in Austria – is poor.
Der bodenbewohnende Milbenkanker Cyphophthalmus duricorius Joseph, 1868 (Fam. Sironidae) galt mehr als 60 Jahre lang als die einzige in Österreich vorkommende Art der Weberknecht-Unterordnung Cyphophthalmi. Neuere Aufsammlungen zeigen jedoch, dass mindestens zwei weitere Cyphophthalmi-Arten in Österreich existieren: 1) Siro cf. crassus Novak & Giribet, 2006 wurde an einer Lokalität in der SW Steiermark nahe der slowenischen Grenze gefunden und repräsentiert eine zweite Gattung von Sironiden in Österreich. 2) Ein weiterer, morphologisch distinkter Sironide („Sironidae gen. et sp. nov.?“) − derzeit noch unbeschrieben und taxonomisch nicht zugeordnet − stammt aus Böden im Grenzgebiet Steiermark-Kärnten. Obwohl bislang kein syntopes Vorkommen belegt ist, können alle drei Arten in einem kleinen Areal von wenigen Quadratkilometern gefunden werden.
The E3 ubiquitin ligase MYCBP2 negatively regulates neuronal growth, synaptogenesis, and synaptic strength. More recently it was shown that MYCBP2 is also involved in receptor and ion channel internalization. We found that mice with a MYCBP2-deficiency in peripheral sensory neurons show prolonged thermal hyperalgesia. Loss of MYCBP2 constitutively activated p38 MAPK and increased expression of several proteins involved in receptor trafficking. Surprisingly, loss of MYCBP2 inhibited internalization of transient receptor potential vanilloid receptor 1 (TRPV1) and prevented desensitization of capsaicin-induced calcium increases. Lack of desensitization, TRPV internalization and prolonged hyperalgesia were reversed by inhibition of p38 MAPK. The effects were TRPV-specific, since neither mustard oil-induced desensitization nor behavioral responses to mechanical stimuli were affected. In summary, we show here for the first time that p38 MAPK activation can inhibit activity-induced ion channel internalization and that MYCBP2 regulates internalization of TRPV1 in peripheral sensory neurons as well as duration of thermal hyperalgesia through p38 MAPK.
Correlations between personality traits and a wide range of sensory thresholds were examined. Participants (N = 124) completed a personality inventory (NEO-FFI) and underwent assessment of olfactory, trigeminal, tactile and gustatory detection thresholds, as well as examination of trigeminal and tactile pain thresholds. Significantly enhanced odor sensitivity in socially agreeable people, significantly enhanced trigeminal sensitivity in neurotic subjects, and a tendency for enhanced pain tolerance in highly conscientious participants was revealed. It is postulated that varied sensory processing may influence an individual's perception of the environment; particularly their perception of socially relevant or potentially dangerous stimuli and thus, varied with personality.
Accumulating evidence indicates that increased generation of reactive oxygen species (ROS) contributes to the development of exaggerated pain hypersensitivity during persistent pain. In the present study, we investigated the antinociceptive efficacy of the antioxidants vitamin C and vitamin E in mouse models of inflammatory and neuropathic pain. We show that systemic administration of a combination of vitamins C and E inhibited the early behavioral responses to formalin injection and the neuropathic pain behavior after peripheral nerve injury, but not the inflammatory pain behavior induced by Complete Freund's Adjuvant. In contrast, vitamin C or vitamin E given alone failed to affect the nociceptive behavior in all tested models. The attenuated neuropathic pain behavior induced by the vitamin C and E combination was paralleled by a reduced p38 phosphorylation in the spinal cord and in dorsal root ganglia, and was also observed after intrathecal injection of the vitamins. Moreover, the vitamin C and E combination ameliorated the allodynia induced by an intrathecally delivered ROS donor. Our results suggest that administration of vitamins C and E in combination may exert synergistic antinociceptive effects, and further indicate that ROS essentially contribute to nociceptive processing in special pain states.
Effect sizes in experimental pain produced by gender, genetic variants and sensitization procedures

Background: Various effects on pain have been reported with respect to their statistical significance, but a standardized measure of effect size has been rarely added. Such a measure would ease comparison of the magnitude of the effects across studies, for example the effect of gender on heat pain with the effect of a genetic variant on pressure pain. Methodology/Principal Findings: Effect sizes on pain thresholds to stimuli consisting of heat, cold, blunt pressure, punctuate pressure and electrical current, administered to 125 subjects, were analyzed for 29 common variants in eight human genes reportedly modulating pain, gender and sensitization procedures using capsaicin or menthol. The genotype explained 0–5.9% of the total interindividual variance in pain thresholds to various stimuli and produced mainly small effects (Cohen's d 0–1.8). The largest effect had the TRPA1 rs13255063T/rs11988795G haplotype explaining >5% of the variance in electrical pain thresholds and conferring lower pain sensitivity to homozygous carriers. Gender produced larger effect sizes than most variant alleles (1–14.8% explained variance, Cohen's d 0.2–0.8), with higher pain sensitivity in women than in men. Sensitization by capsaicin or menthol explained up to 63% of the total variance (4.7–62.8%) and produced largest effects according to Cohen's d (0.4–2.6), especially heat sensitization by capsaicin (Cohen's d = 2.6). Conclusions: Sensitization, gender and genetic variants produce effects on pain in the mentioned order of effect sizes. The present report may provide a basis for comparative discussions of factors influencing pain.
Meeting Abstract : 10. Deutscher Kongress für Versorgungsforschung, 18. GAA-Jahrestagung. Deutsches Netzwerk Versorgungsforschung e. V. ; Gesellschaft für Arzneimittelanwendungsforschung und Arzneimittelepidemiologie e. V. 20.-22.10.2011, Köln
Hintergrund: Multimedikation als Folge von Multimorbidität ist ein zentrales Problem der Hausarztpraxis und erhöht das Risiko für unangemessene Arzneimittel-Verordnungen (VO). Um die Medikation bei älteren, multimorbiden Patienten zu optimieren und zu priorisieren, wurde eine computergestützte, durch Medizinische Fachangestellte (MFA) assistierte, komplexe Intervention (checklistengestütztes Vorbereitungsgespräch sowie Überprüfung eingenommener Medikamente durch MFA, Einsatz des web-basierten ArzneimittelinformationsDienstes AiD, spezifisches Arzt-Patienten-Gespräch) entwickelt und in einer 12-monatigen Pilotstudie auf Machbarkeit getestet. Ein auf 9 Items reduzierter MAI [1] wurde eingesetzt, um dessen Eignung als potentielles primäres Outcome der Hauptstudie zu prüfen.
Material und Methoden: In die Pilotstudie in 20 Hausarztpraxen mit Cluster-Randomisation auf Praxisebene in Kontrollgruppe (Regelversorgung b. empfohlenem Standard) vs. Interventionsgruppe (komplexe Intervention b. empfohlenem Standard) wurden 5 Pat./Praxis eingeschlossen (≥65 Jahre, ≥3 chron. Erkrankungen, ≥5 Dauermedikamente, MMSE ≥26, Lebenserwartung ≥6 Monate). Zur Bewertung des MAI wurden an Baseline (T0), 6 Wo. (T1) & 3 Mon. (T2) nach Intervention erhoben: VO, Diagnosen, Natrium, Kalium & Kreatinin i.S., Größe, Gewicht, Geschlecht, Cumulative Illness Rating Scale (CIRS) [2] durch die Hausarztpraxis; Symptome für unerwünschte Arzneimittelwirkungen im Patienten-Telefoninterview.
Für den MAI wurde die Angemessenheit jeder VO in den 9 Kategorien Indikation, Effektivität, Dosierung, korrekter & praktikabler Applikationsweg, Arzneimittelwechselwirkung, Drug-disease-Interaktion, Doppelverordnung, Anwendungsdauer 3-stufig bewertet (1 = korrekt - 3 = unkorrekt) und für die Auswertung auf Patientenebene summiert. Die Bewertung erfolgte ohne Kenntnis der Gruppenzugehörigkeit. Deskriptive Statistiken und Reliabilitätsanalysen, ungewichtete Auswertung und Gewichtung n. Bregnhoj [3].
Ergebnisse: Es wurden N=100 Patienten in die Studie eingeschlossen, im Mittel 76 Jahre (Standardabweichung, SD 6; Range, R: 64-93) , 52% Frauen, durchschnittlich 9 VO/Pat. (SD 2; R 4-16), mittlerer CIRS-Score 10 (SD 4; R 0-23). Basierend auf N=851 VO (100 Pat.) zu T0 betrug der Reliabilitätskoeffizient (RK, Cronbachs Alpha) der ungewichteten 9 Items 0,70. Items 1-5 wiesen akzeptable Trennschärfen auf (0,52-0,64), die der Items 6, 7 & 9 fielen mit 0,21-0,29 niedriger aus, die des Item 8 betrug 0,06. Auf der Basis der 9 gewichteten Items fiel die interne Konsistenz des MAI erwartet höher aus (0,75). Die Reliabilitätsanalysen auf VO-Ebene zeigten einen RK von 0,67 (ungewichtet) vs. 0,75 (gewichtet), die Trennschärfen waren vergleichbar. Zur Zwischenauswertung betrug der MAI (T1-T0) in der Interventionsgruppe (5 Praxen, 24 Pat.) -0,9 (SD 5,6), in der Kontrollgruppe (7 Praxen, 35 Pat.) -0,5 (SD 4,9); die Differenz zwischen beiden Gruppen Mi–Mk -0,4 [95% Konfidenzintervall: -3,4;2,6].
Schlussfolgerung: Der MAI ist als potentielles primäres Outcome in der Hauptstudie geeignet: wenige fehlende Werte, Darstellung von Unterschieden prä-post und zwischen den Gruppen, akzeptable interne Konsistenz. Der niedrige Trennschärfekoeffizient des Items 8 weist darauf hin, dass dieses Item nicht mit dem Gesamt-Skalenwert korreliert, auch die Items 6, 7 und 9 korrelieren wesentlich schwächer mit dem Gesamt-Skalenwert als die Items 1 bis 5. Eine Wichtung z.B. der Items 2, 5, 6 und 9 könnte erwogen werden, um den Fokus der Intervention in der Hauptzielgröße angemessen abzubilden.
Background: MicroRNA-21 (miR-21) is up-regulated in tumor tissue of patients with malignant diseases, including hepatocellular carcinoma (HCC). Elevated concentrations of miR-21 have also been found in sera or plasma from patients with malignancies, rendering it an interesting candidate as serum/plasma marker for malignancies. Here we correlated serum miR-21 levels with clinical parameters in patients with different stages of chronic hepatitis C virus infection (CHC) and CHC-associated HCC.
Methodology/Principal Findings: 62 CHC patients, 29 patients with CHC and HCC and 19 healthy controls were prospectively enrolled. RNA was extracted from the sera and miR-21 as well as miR-16 levels were analyzed by quantitative real-time PCR; miR-21 levels (normalized by miR-16) were correlated with standard liver parameters, histological grading and staging of CHC. The data show that serum levels of miR-21 were elevated in patients with CHC compared to healthy controls (P<0.001); there was no difference between serum miR-21 in patients with CHC and CHC-associated HCC. Serum miR-21 levels correlated with histological activity index (HAI) in the liver (r = −0.494, P = 0.00002), alanine aminotransferase (ALT) (r = −0.309, P = 0.007), aspartate aminotransferase (r = −0.495, P = 0.000007), bilirubin (r = −0.362, P = 0.002), international normalized ratio (r = −0.338, P = 0.034) and γ-glutamyltransferase (r = −0.244, P = 0.034). Multivariate analysis revealed that ALT and miR-21 serum levels were independently associated with HAI. At a cut-off dCT of 1.96, miR-21 discriminated between minimal and mild-severe necroinflammation (AUC = 0.758) with a sensitivity of 53.3% and a specificity of 95.2%.
Conclusions/Significance: The serum miR-21 level is a marker for necroinflammatory activity, but does not differ between patients with HCV and HCV-induced HCC.
Background: Threonine Aspartase 1 (Taspase1) mediates cleavage of the mixed lineage leukemia (MLL) protein and leukemia provoking MLL-fusions. In contrast to other proteases, the understanding of Taspase1's (patho)biological relevance and function is limited, since neither small molecule inhibitors nor cell based functional assays for Taspase1 are currently available. Methodology/Findings: Efficient cell-based assays to probe Taspase1 function in vivo are presented here. These are composed of glutathione S-transferase, autofluorescent protein variants, Taspase1 cleavage sites and rational combinations of nuclear import and export signals. The biosensors localize predominantly to the cytoplasm, whereas expression of biologically active Taspase1 but not of inactive Taspase1 mutants or of the protease Caspase3 triggers their proteolytic cleavage and nuclear accumulation. Compared to in vitro assays using recombinant components the in vivo assay was highly efficient. Employing an optimized nuclear translocation algorithm, the triple-color assay could be adapted to a high-throughput microscopy platform (Z'factor = 0.63). Automated high-content data analysis was used to screen a focused compound library, selected by an in silico pharmacophor screening approach, as well as a collection of fungal extracts. Screening identified two compounds, N-[2-[(4-amino-6-oxo-3H-pyrimidin-2-yl)sulfanyl]ethyl]benzenesulfonamideand 2-benzyltriazole-4,5-dicarboxylic acid, which partially inhibited Taspase1 cleavage in living cells. Additionally, the assay was exploited to probe endogenous Taspase1 in solid tumor cell models and to identify an improved consensus sequence for efficient Taspase1 cleavage. This allowed the in silico identification of novel putative Taspase1 targets. Those include the FERM Domain-Containing Protein 4B, the Tyrosine-Protein Phosphatase Zeta, and DNA Polymerase Zeta. Cleavage site recognition and proteolytic processing of these substrates were verified in the context of the biosensor. Conclusions: The assay not only allows to genetically probe Taspase1 structure function in vivo, but is also applicable for high-content screening to identify Taspase1 inhibitors. Such tools will provide novel insights into Taspase1's function and its potential therapeutic relevance.
Nerve injury leads to sensitization mechanisms in the peripheral and central nervous system which involve transcriptional and post-transcriptional modifications in sensory nerves. To assess protein regulations in the spinal cord after injury of the sciatic nerve in the Spared Nerve Injury model (SNI) we performed a proteomic analysis using 2D-difference gel electrophoresis (DIGE) technology. Among approximately 2300 protein spots separated on each gel we detected 55 significantly regulated proteins after SNI whereof 41 were successfully identified by MALDI-TOF MS. Out of the proteins which were regulated in the DIGE analyses after SNI we focused on the carboxypeptidase A inhibitor latexin because protease dysfunctions contribute to the development of neuropathic pain. Latexin protein expression was reduced after SNI which could be confirmed by Western Blot analysis, quantitative RT-PCR and in-situ hybridisation. The decrease of latexin was associated with an increase of the activity of carboxypeptidase A indicating that the balance between latexin and carboxypeptidase A was impaired in the spinal cord after peripheral nerve injury due to a loss of latexin expression in spinal cord neurons. This may contribute to the development of cold allodynia because normalization of neuronal latexin expression in the spinal cord by AAV-mediated latexin transduction or administration of a small molecule carboxypeptidase A inhibitor significantly reduced acetone-evoked nociceptive behavior after SNI. Our results show the usefulness of proteomics as a screening tool to identify novel mechanisms of nerve injury evoked hypernociception and suggest that carboxypeptidase A inhibition might be useful to reduce cold allodynia.
Background: The treatment of high-risk neuroblastoma patients consists of multimodal induction therapy to achieve remission followed by consolidation therapy to prevent relapses. However, the type of consolidation therapy is still discussed controversial. We applied metronomic chemotherapy in the prospective NB90 trial and monoclonal anti-GD2-antibody (MAB) ch14.18 in the NB97 trial. Here, we present the long term outcome data of the patient cohort. Methods: A total of 334 stage 4 neuroblastoma patients one year or older were included. All patients successfully completed the induction therapy. In the NB90 trial, 99 patients received at least one cycle of the oral maintenance chemotherapy (NB90 MT, 12 alternating cycles of oral melphalan/etoposide and vincristine/cyclophosphamide). In the NB97 trial, 166 patients commenced the MAB ch14.18 consolidation therapy (six cycles over 12 months). Patients who received no maintenance therapy according to the NB90 protocol or by refusal in NB97 (n = 69) served as controls. Results: The median observation time was 11.11 years. The nine-year event-free survival rates were 41 ± 4%, 31 ± 5%, and 32 ± 6% for MAB ch14.18, NB90 MT, and no consolidation, respectively (p = 0.098). In contrast to earlier reports, MAB ch14.18 treatment improved the long-term outcome compared to no additional therapy (p = 0.038). The overall survival was better in the MAB ch14.18-treated group (9-y-OS 46 ± 4%) compared to NB90 MT (34 ± 5%, p = 0.026) and to no consolidation (35 ± 6%, p = 0.019). Multivariable Cox regression analysis revealed ch14.18 consolidation to improve outcome compared to no consolidation, however, no difference between NB90 MT and MAB ch14.18-treated patients was found. Conclusions: Follow-up analysis of the patient cohort indicated that immunotherapy with MAB ch14.18 may prevent late relapses. Finally, metronomic oral maintenance chemotherapy also appeared effective.
A catalogue of aphidiine parasitoids (Hymenoptera: Braconidae: Aphidiinae) associated with various aphids species occurring in India was compiled. The present catalogue with 125 species under 22 genera has been further reinforced with not only all the latest taxonomic changes but also host names, host plants, distribution in India etc.

Bericht zur Tagung 'From Arthouse to Grindhouse - and back? Wechselbeziehungen zwischen Hoch- und Populärkultur', Abteilung für Vergleichende Literaturwissenschaft der Universität Wien, Societa - Forum für Ethik, Kunst und Recht, Filmarchiv Austria, 5. bis 7. Mai 2011
Die von Keyvan Sarkhosh (Wien) und Paul Ferstl (Wien) organisierte Tagung, die im Studiokino des Filmarchivs Austria stattfand, widmete sich in dreizehn Vorträgen diesen komplexen, sprach- und medienübergreifenden Wechselbeziehungen von 'Arthouse' und 'Grindhouse' und verband die inhaltliche Thematik mit dem methodischen Ziel einer verstärkten Theoretisierung bei der Betrachtung populärkultureller Phänomene.
Medizin gehört auch nach über 40 Jahren zu den zulassungsbeschränkten Fächern; die
Zulassung nach Leistung (Abiturnote) und Wartezeit befriedigt dabei nicht vollständig.
Die Eignung alternativer Kriterien für die Studienzulassung wurde bisher kaum
untersucht; angesichts der je nach Bundesland und Leistungsfächern heterogenen
Abituranforderungen wurden bisher auch keine Medizin-spezifischen Kriterien für die
Eignung als Zulassungskriterium untersucht, weder als Teilleistungen des Abiturs noch
als extracurriculare Aktivitäten.
Die vorliegende Studie untersucht die Bedeutung naturwissenschaftlicher Vorkenntnisse
für das Medizinstudium. Hierzu wurden das aktive Wissen in den Fächern Biologie,
Chemie, Physik und Mathematik in einer schriftlichen Klausur mit 40 offenen Fragen
an 3030 Studenten überprüft, was etwa 40% der Studienanfänger des Semesters an 14
Universitäten in Deutschland entsprach. Die Klausur wurde jeweils in den ersten vier
Wochen ihres ersten vorklinischen Semesters im Wintersemester 2004/2005
geschrieben; das Schwierigkeitsniveau der Fragen entsprach dem Lehrstoff der
Klassenstufen 10 bis 12 des Gymnasiums.
Im Durchschnitt beantworteten die Studienanfänger dabei 14,34 der 40 Fragen richtig
(36%). Aufgeteilt in die vier naturwissenschaftlichen Fächer zeigte sich, dass in den
Fächern Biologie und Mathematik jeweils über 40% der Fragen richtig beantwortet
wurden, während der Anteil der richtigen Antworten in den Fächern Chemie und Physik
mit 22% beziehungsweise 19% deutlich niedriger lag.
Die Studienanfänger in der Bundesrepublik Deutschland zeigten erhebliche
Wissenslücken auf. Im Weiteren wird in der vorliegenden Studie der Zusammenhang
zwischen den naturwissenschaftlichen Abiturnoten und -fächern der Studenten, den
naturwissenschaftlichen Kenntnissen (gemäß Klausur) und dem Studienerfolg,
gemessen an den Physikumsnoten, untersucht.
Anzustreben wäre eine bessere Vorbereitung der Studienanwärter in grundlegenden
naturwissenschaftlichen Kenntnissen, entweder in den Gymnasien oder in den ersten
vorklinischen Semestern. Dies könnte durch Medizin-spezifische Aufnahmetests vor Studienbeginn oder durch zusätzlich angebotene, freiwillige oder verpflichtende Kurse
in den naturwissenschaftlichen Fächern erfolgen, um den späteren Studienerfolg zu
gewährleisten beziehungsweise zu verbessern.
According to the World Health Organization (WHO) bacterial resistance to antibiotic drug therapy is emerging as a major public health problem around the world. Infectious diseases seriously threaten the health and economy of all countries. Hence, the preservation of the effectiveness of antibiotics is a world wide priority. The key to preserving the power of antibiotics lies in maintaining their diversity. Many microorganisms are capable of producing these bioactive products, the so called antibiotics. Specifically in microorganisms, polyketide synthases (PKS) and non-ribosomal peptide synthases (NRPS) produce these natural bioactive compounds. Besides being used as antibiotics these non-ribosomal peptides and polyketides display an even broader spectrum of biological activities, e.g. as antivirals, immunosuppressants or in antitumor therapy. The wide functional spectrum of the peptides and ketides is due to their structural diversity. Mostly they are cyclic or branched cyclic compounds, containing non-proteinogenic amino acids, small heterocyclic rings and other unusual modifications such as epimerization, methylation, N‐formylation or heterocyclization. It is has been shown that these modifications are important for biological activity, but little is known about their biosynthetic origin.
PKS and NRPS are multidomain protein assembly lines which function by sequentially elongating a growing polyketide or peptide chain by incorporating acyl units or amino acids, respectively. The growing product is attached via a thioester linkage to the 4’-phosphopantetheine (4’-Ppant) arm of a holo acyl carrier protein (ACP) in PKSs or holo peptidyl carrier protein (PCP) in NRPSs and is passed from one module to another along the chain of reaction centers. The modular arrangement makes PKS and NRPS systems an interesting target for protein engineering. More than 200 novel polyketide compounds have already been created by module swapping, gene deletion or other specific manipulations. Unfortunately, however, engineered PKS often fail to produce significant amounts of the desired products. Structural studies may faciliate yield improvement from engineered systems by providing a more complete understanding of the interface between the different domains. While some information about domain-domain interactions, involving the most common enzymatic modules, ketosynthase and acyltransferase, is starting to emerge, little is known about the interaction of ACP domains with other modifying enzymes such as methyltransferases, epimerases or halogenases.
To further improve the understanding of domain-domain interactions this work focuses on the curacin A assembly line. Curacin A, which exhibits anti-mitotic activity, is from the marine cyanobacterium Lyngbya majuscula. This outstanding natural product contains a cyclopropane ring, a thiazoline ring, an internal cis double bond and a terminal alkene. The biosynthesis of curacin A is performed by a 2.2 Mega Dalton (MDa) hybrid PKS-NRPS cluster. A 10-enzyme assembly catalyzes the formation of the cyclopropane moiety as the first building block of the final product. Interestingly, for these enzymes the substrate is presented by an unusual cluster of three consecutive ACPs (ACPI,II,III). Little is known about the function of multiple ACPs which are supposed to increase the overall flux for enhanced production of secondary metabolites.
The first task in this work was to elucidate the structural effect of the triplet ACP repetition by nuclear magnetic resonance (NMR). The initial data show that the excised ACPI, ACPII or ACPIII proteins resulted in [15N, 1H]-TROSY spectra with strong chemical shift perturbations (CSPs), suggesting an effect on the structure. The triplet ACP domains display a high sequence identity (93- 100%) making structural investigation using usual NMR techniques due to high peak overlap impossible. To enable the investigation of the triplet ACP in its native composition we developed a powerful method, the three fragment ligation. Segmental labeling allows incorporating isotopes into one single domain in its multidomain context. As a result we could prepare the triplet ACP with only one domain isotopically labeled and therefore assign the full length protein. In this way our method paved the way to study the structural effects of the triplet ACP repetition. We could show unexpectedly, that, despite the fact that the triplet repeat of CurA ACPI,II,III has a synergistic effect in the biosynthesis of CurA, the domains are structurally independent.
In the second part of this work, we studied the structure of the isolated ACPI domain. Our results show that the CurA ACPI undergoes no major conformational changes upon activation via phosphopantetheinylation and therefore contradicts the conformational switching model which has been proposed for PCPs. Further we report the NMR solution structures of holo-ACPI and 3-hydroxyl-3-methylglutaryl (HMG)-ACPI. Data obtained from filtered nuclear overhauser effect (NOE) experiments indicate that the substrate HMG is not sequestered but presented on the ACP surface.
In the third part of this work we focussed on the protein-protein interactions of the isolated ACPI with its cognate interaction partners. We were especially interested in the interaction with the halogenase (Cur Hal), the first enzyme within the curacin A sub-cluster, acting on the initial hydroxyl-methyl-glutaryl (HMG) attached to ACPI. Primarily we studied the interaction using NMR titration and fluorescence anisotropy measurements. Surprisingly no complex between ACPI and Cur Hal could be detected. The combination of an activity assay using matrix-assisted laser desorption/ionization (MALDI) mass spectroscopy and mutational analysis revealed several amino acids of ACPI that strongly decrease the activity of CurA Hal. Mapping these mutations according to their effect on the Cur Hal activity onto the structure of HMG-ACPI displays that these amino acids surround the substrate and form a consecutive surface. These results suggest that this surface is important for Cur Hal recognition and selectivity. Our research presented herein is an excellent example for protein-protein interactions in PKS systems underlying a specific recognition process.
Die Familie der Proteorhodopsine (PR) besteht aus Hunderten von PR Molekülen, die unter Lichteinwirkung Protonen pumpen und somit eine bedeutende Rolle für die Energiegewinnung spielen könnten. Da der pKa Wert des Proton Akzeptors der Schiff‘schen Base (SB) (~7.2) dem pH Wertes der Ozeane (~7.9) ähnelt, wird auch über eine regulatorische Funktion spekuliert. Wird in Erwägung gezogen, dass 24 000 PR Moleküle pro SAR86 Zelle vorhanden sind (Beja et al. 2001) und dass 13% der Bakterien der Meeresoberfläche PR besitzen (Sabehi et al. 2005) liefert dieses Protein wahrscheinlich einen bedeutenden Energiebeitrag neben der Photosynthese. Einblicke in den Mechanismus der Energieumwandlung erfordern sowohl die Untersuchung des Chromophores, welches die Lichtenergie absorbiert als auch der Struktur des Apoproteins, das durch die Generierung eines Protonengradienten zur Energiegewinnung beiträgt. Der Fokus der Doktorarbeit liegt auf dem Chromophor und seiner Umgebung. Eine erste Charakterisierung der SB und des Retinals erfolgt durch UV/VIS und NMR Messungen (Pfleger et al. 2008). Die 13C chemische Verschiebungen von 10,11-13C2 Retinal und die 15N chemische Verschiebung der protonierten SB, gebildet durch K231, zeigt eindeutig, dass im Grundzustand nur eine Konformation der Retinals, all-trans, vorliegt. Die 15N chemische Verschiebung weist außerdem auf eine starke Wechselwirkung der SB mit ihren Gegenionen hin. Desweiteren kann durch Messungen der 15N chemischen Verschiebung der SB bei verschiedenen pH Werten der pKa Wert der SB abgeschätzt werden, auf ~12. Diese Stabilisierung der positiv geladenen protonierten Form der SB weist auf die Existenz eines Wasserclusters hin, das durch die hohe Dielektrizitätskonstante die protonierte Form der SB stabilisieren könnte. Um zu überprüfen, ob Wasser an der SB gebunden ist, wird ein sogenanntes 15N-1H HETCOR Experiment durchgeführt. Der Bereich der 15N chemischen Verschiebung der SB korreliert mit einer Protonenresonanz bei ~5 ppm, welche im Bereich einer Wasserresonanz liegt und die durch D2O austauschbar ist. Dies indiziert eine wichtige Bedeutung von Wasser in der Nähe der SB für die Funktion von PR. Der Einfluss von Mutationen des Histidins H75 und des Aspartats D97 auf die 15N chemische Verschiebung der SB sowie die Auswirkung von Histidinmutationen auf das Chromophor deuten eine direkte Wechselwirkung von Aspartat 97 und der SB an, nicht aber eine direkte Wechselwirkung von H75 und der SB. Neben dem Chromophor ist außerdem das Signalpeptid Gegenstand der Untersuchung der Doktorarbeit. Motivation für die Untersuchung war die Inhomogenität der Proben, die im Zusammenhang mit ungleich prozessiertem PR stehen könnten. Ein zweiter Teil beschäftigt sich mit neuen Konzepten der Datenaufnahme, da das S/R in der Festkörper NMR ein limitierender Faktor darstellt. Diese beinhalten Verstärkung der Relaxation (RELOAD) sowie die Refokussierung von T2 bei Verwendung eines Prozessierungsschrittes, der „half echo alternating transformation“ (HEAT).
Diese Arbeit benutzt mikrometeorologische, pflanzenökologische und bodenhydrologische Messungen als Mittel zum Prozessverständnis. Der langfristige Flussmessstandort Ankerstation Tharandter Wald (von 120 jährigen Fichten dominiert) zeigt die große Dynamik der Landoberflächen-Atmosphären-Wechselwirkungen wie auch ihre Klimaeffekte auf die Verteilung der turbulenten Wärmeströme, die Kohlenstoffsequestrierung und die Evapotranspiration (ET). Klimawerte, Phänologie und Flüsse unterstützen die Einteilung des Jahres in eine ‚aktive Phase’ (April–September) und eine ‚Ruhephase’ (Oktober– März): Kohlenstoffsequestrierung, zur Verfügung stehende Energie (Strahlungsbilanz) und fühlbarer Wärmestrom sind in der Ruhephase praktisch vernachlässigbar. Nur ET zeigt einen signifikanten Beitrag zur Jahresbilanz (25 % der aktiven Phase) aus der Interzeption (Evaporation von benetzten Nadeln), die vom fühlbaren Wärmestrom aus der Atmosphäre angetrieben wird. Die zwischenjährliche Variation der Flüsse ist im Allgemeinen klein (z. B. 500–650 gC m-2 yr -1) C-Aufnahme), selbst mit dem starken Dürrejahr 2003 (400 gC m-2) oder dem Effekt der Durchforstung 2002. Verglichen mit der Buche erreicht die Fichte – zumindest in der aktiven Periode – ähnliche Werte von ET aber niedrigere bei der C-Aufnahme. Die Kronentraufe beträgt bei der Fichte nur ca. 55 % des Niederschlages, bei der Buche summieren sich ca. 40 % Kronentraufe und knapp 25 % Stammabfluss zu etwa 65 % Bestandesniederschlag. Dieser Unterschied erklärt möglicherweise die im Allgemeinen höhere Bodenfeuchte am Buchenstandort. Als Resultat aus dieser Arbeit werden Modelle mit ausreichender Komplexität empfohlen, welche Bestandesstruktur und Phänophasen berücksichtigen. Das ist eine Voraussetzung für eine bessere Berücksichtigung von Wäldern mit ihren Landoberflächen-
Atmosphären-Wechselwirkungen, z. B. in Klimamodellen.
The enzyme quinol:fumarate reductase (QFR) from the anaerobic epsilon-proteobacterium Wolinella succinogenes is a membrane protein complex that couples the catalysis of the oxidation of menaquinol to menaquinone to that of the reduction of fumarate to succinate. This is the terminal step in fumarate respiration, a form of anaerobic respiration in which oxygen is replaced by fumarate as the terminal electron acceptor in many anaerobic microorganisms. In QFR, both the heme groups (low-potential distal and high-potential proximal heme b group in transmembrane subunit C) are part of the electron transport chain between the two catalytic sites of the redox enzyme. Although the reduction of fumarate by menaquinol is exergonic, it is not exergonic enough to support the generation of a transmembrane electrochemical proton potential delta p. Evidence has previously shown that this reaction is catalysed by a novel mechanism, involving the facilitation of transmembrane electron transfer by transmembrane proton transfer via an essential compensatory transmembrane proton transfer pathway ("E-pathway") which is inactive in the oxidized state of the enzyme. The two key constitutents of the the pathway are the amino acid residue Glu C180 of the transmembrane helix V (located in subunit C) and the ring C propionate of the distal heme bD. The aim of the project was to obtain, by employing a combination of time-resolved as well as static spectroscopic approaches, a detailed insight of the transmembrane electron coupled proton transfer mechanism. Minute changes in both the oxidized and reduced states of a redox protein system can be selectively and sensitively monitored by static Fourier Transformed Infrared (FTIR) difference spectroscopy. The technique employed in this context, electrochemically induced FTIR difference spectroscopy, is complemented by computer-based electrostatic calculations. In order to elucidate the catalytic mechanism of the important reactions in QFR, it is necessary to investigate these in a time-resolved manner. Rapid scan FTIR difference spectroscopy is a suitable technique that allows the course of the reaction to be monitored in a time dependent fashion. The techniques employed in this context are time-resolved (tr-FTIR) and transient absorption spectroscopy. In the following, the details of individual sub-projects are discussed in brief. ...

Grundlegend für eine solche Forstwirtschaft ist unter anderem die räumlich-differenzierte Erfassung und Bewertung wasserhaushaltsbezogener Standortsmerkmale. Denn erst die Kenntnis der Dynamik des pflanzenverfügbaren Bodenwasserangebotes oder auch eines möglichen Überschusses in Form von Stauwasser erlaubt eine standortsgerechte Baumartenwahl als Voraussetzung für eine nachhaltige Waldbewirtschaftung.
Mitochondrial complex I, the largest and most complicated proton pump of the respiratory chain, links the electron transfer from NADH to ubiquinone to the pumping of four protons from the matrix into the intermembrane space. In humans, defects in complex I are involved in a wide range of degenerative disorders. Recent progress in the X-ray structural analysis of prokaryotic and eukaryotic complex I confirmed that the redox reactions are confined entirely to the hydrophilic peripheral arm of the L-shaped molecule and take place at a remarkable distance from the membrane domain. While this clearly implies that the proton pumping within the membrane arm of complex I is driven indirectly via long-range conformational coupling, the molecular mechanism and the number, identity, and localization of the pump-sites remains unclear. Here, we report that upon deletion of the gene for a small accessory subunit of the Yarrowia complex I, a stable subcomplex (nb8m delta) is formed that lacks the distal part of the membrane domain as revealed by single particle analysis. The analysis of the subunit composition of holo and subcomplex by three complementary proteomic approaches revealed that two (ND4 and ND5) of the three subunits with homology to bacterial Mrp-type Na+/H+ antiporters that have been discussed as prime candidates for harbouring the proton pumps were missing in nb8m delta. Nevertheless, nb8m delta still pumps protons at half the stoichiometry of the complete enzyme. Our results provide evidence that the membrane arm of complex I harbours two functionally distinct pump modules that are connected in series by the long helical transmission element recently identified by X-ray structural analysis.
Die Transpiration eines Fichten– und Buchenbestandes unter Bodentrockenheit im Tharandter Wald

Für die Bewertung des Standortwasserhaushaltes sind die Anforderungen der Bestockung unter sich ändernden Umweltbedingungen mit zu berücksichtigen. Von den unterschiedlichen Wasserhaushaltskomponenten gibt die Bestandestranspiration die physiologische Reaktion der Bäume wider und steht somit auch dem Kohlenstoffhaushalt am nächsten. Eine regulierende und somit kritische Größe stellt die Bestandesleitfähigkeit dar, die zwischen Wasser- und Kohlenstoffhaushalt vermittelt. Als Indikator für diese Regulation wird das Verhältnis von Kohlenstoffaufnahme zu Wasserabgabe, die sog. Wassernutzungseffizienz, herangezogen. An einem Fichten- und Buchenstandort wurde mittels Xylemsaftflussmessungen die Bestandestranspiration (Ec) und -leitfähigkeit (gc) bestimmt. Von Mai bis Oktober 2006 lag Ec am Fichtenstandort (161 mm Saison-1) deutlich unter Ec von Buche mit 182 mm Saison-1 während die Gesamtjahresbilanz ähnlich war. Dies stand in Beziehung mit einer Phase sommerlicher Bodentrockenheit, die sich am Fichtenstandort deutlicher als am Buchenstandort auswirkte. Im Gegensatz zu Buche war während der Bodentrockenheit die Bestandesleitfähigkeit von Fichte, unabhängig von atmosphärischen Bedingungen, auf minimale Werte reduziert und die Wassernutzungseffizienz erhöht. Aus den Befunden kann gefolgert werden, dass Standortverhältnisse und Baumarten zu messbaren Unterschieden im Standortswasserhaushalt führen, die auch in Wasserhaushaltsmodellen darstellbar sein sollten. Die vorliegenden Messdaten sind grundlegend, um transpirationsbezogene Indikatoren von Wasserhaushaltsmodellen zu bewerten.
Die forstliche Standortklassifikation ist bundesweit uneinheitlich und bezüglich der Bewertung des Gesamtwasserhaushalts meist subjektiv. Um eine Vereinheitlichung zu erreichen, wird eine modellbasierte, objektive Klassifikation angestrebt, die neben der reliefbasierten meteorologischen Variabilität auch Bodenform und Bestockung einbezieht. In diesem Artikel werden die Ergebnisse vergleichender BROOK 90-Simulationen, die den Effekt unterschiedlicher Böden, Baumarten und Ausrichtungen im Gelände untersuchen, dargestellt und diskutiert. Der Schwerpunkt liegt dabei auf der Betrachtung von Wasserverfügbarkeit. Es wurden Parametrisierungen für vier verschiedene Bodenformen mit den Bestockungen Buche, Eiche, Fichte und Kiefer verwendet. Die reliefbedingten meteorologischen Standortcharakteristiken richten sich nach den lokalen Gegebenheiten des Testgebiets Tharandter Wald. Für das Konzept der Darstellung von Unterschieden hinsichtlich des Standortswasserhaushaltes wurden hierfür im Modell implementierte Stressindikatoren verwendet.
Es zeigt sich, daß in Trockenjahren die Laubbäume höhere Stressindikatoren erreichen. Gleichfalls treten bei Böden mit geringerer Wasserretention erwartungsgemäß mehr Stress-tage auf. Bestockung und Bodeneigenschaften haben einen Einfluß auf Wasserknappheit in der Größenordnung wie die zur forstlichen Standortklassifikation verwendeten morphologischen Geländeeigenschaften.
Auf der Basis des forsthydrologischen Modells BROOK 90 wurde ein Modellsystem zur Berechnung und Beurteilung des Standortswasserhaushaltes in Abhängigkeit von Klima, Relief, Boden und Bestockung entwickelt. Implementierte Indikatoren zur Ausschöpfung des Bodenwasservorrates, der Einschränkung der Transpiration und des Auftretens von Staunässe erlauben eine Visualisierung der Modellergebnisse in Form neuartiger Standortskarten. Basierend auf der Kenntnis der Unterschreitungshäufigkeiten von Schwellenwerten dieser Indikatoren wurde ein fünfstufiger Bewertungsrahmen für den Wasserhaushalt aufgestellt. Das Modell wurde im Tharandter Wald für ein Gebiet mit variierenden Böden und unterschiedlicher Bestockung (Buche, Eiche, Fichte und Kiefer) getestet. Die Ergebnisse zeigen, dass diese Vorgehensweise eine differenzierte Informationsgrundlage für die forstliche Planung liefert. Beispielsweise können Entscheidungen zu Baumartenwahl und Bestandesstruktur abgeleitet werden. Forschungsbedarf besteht vor allem in der Verknüpfung der im Modell implementierten Indikatoren des Wasserhaushaltes mit Wachstumsparametern und physiologisch definierten Schwellenwerten.

Ist ein Cyberangriff demnach in seiner Schwere äquivalent zu einem Angriff in der realen Welt, so muss der Angreifer mit Gegenschlägen mit konventionellen Waffen rechnen...
Am vergangenen Freitag gab es im Bundestag eine Debatte über Netzneutralität. Diese konnte ich leider nicht live verfolgen, weil parallel die Arbeitsgruppe Urheberrecht der Enquete-Kommission tagte. Heise berichtet aber über die Debatte und ich gehe hier nur mal auf die dort zitierten Beiträge und Argumente ein. Besonders eingeschlagen hat das plakative Statement der FDP "Netzneutralität ist Internet-Sozialismus". ...
Mit dem wachsenden Interesse für die Vorgänge des Lebendigen im 19. Jahrhundert rückten auch subjektive Erfahrungen in den Bereich lebenswissenschaftlicher Forschung. Damit wurde nicht zuletzt die Frage nach der adäquaten Perspektive virulent, von der aus sich ein Wissen über diese Innerlichkeiten generieren ließe. Das Buch nimmt in drei Fallstudien das neurologische Selbstexperiment Henry Heads, Jacques-Joseph Moreau de Tours' psychiatrische Versuche, den Wahnsinn mit Haschisch zu modellieren, und Benjamin Paul Bloods philosophisch ambitionierten Lachgaskonsum in den Blick, deren Protagonisten sich allesamt dafür entschieden die Innenperspektive einzunehmen und an sich selbst zu experimentieren.
Integrin-mediated cell-matrix interaction in physiological and pathological blood vessel formation

Physiological as well as pathological blood vessel formation are fundamentally dependent on cell-matrix interaction. Integrins, a family of major cell adhesion receptors, play a pivotal role in development, maintenance, and remodeling of the vasculature. Cell migration, invasion, and remodeling of the extracellular matrix (ECM) are integrin-regulated processes, and the expression of certain integrins also correlates with tumor progression. Recent advances in the understanding of how integrins are involved in the regulation of blood vessel formation and remodeling during tumor progression are highlighted. The increasing knowledge of integrin function at the molecular level, together with the growing repertoire of integrin inhibitors which allow their selective pharmacological manipulation, makes integrins suited as potential diagnostic markers and therapeutic targets.

Background
Cytochrome-P450 (CYP450) epoxygenases metabolise arachidonic acid (AA) into four different biologically active epoxyeicosatrienoic acid (EET) regioisomers. Three of the EETs (i.e., 8,9-, 11,12- and 14,15-EET) are rapidly hydrolysed by the enzyme soluble epoxide hydrolase (sEH). Here, we investigated the role of sEH in nociceptive processing during peripheral inflammation.
Results
In dorsal root ganglia (DRG), we found that sEH is expressed in medium and large diameter neurofilament 200-positive neurons. Isolated DRG-neurons from sEH-/- mice showed higher EET and lower DHET levels. Upon AA stimulation, the largest changes in EET levels occurred in culture media, indicating both that cell associated EET concentrations quickly reach saturation and EET-hydrolyzing activity mostly effects extracellular EET signaling. In vivo, DRGs from sEH-deficient mice exhibited elevated 8,9-, 11,12- and 14,15-EET-levels. Interestingly, EET levels did not increase at the site of zymosan-induced inflammation. Cellular imaging experiments revealed direct calcium flux responses to 8,9-EET in a subpopulation of nociceptors. In addition, 8,9-EET sensitized AITC-induced calcium increases in DRG neurons and AITC-induced calcitonin gene related peptide (CGRP) release from sciatic nerve axons, indicating that 8,9-EET sensitizes TRPA1-expressing neurons, which are known to contribute to mechanical hyperalgesia. Supporting this, sEH-/- mice showed increased nociceptive responses to mechanical stimulation during zymosan-induced inflammation and 8,9-EET injection reduced mechanical thresholds in naive mice.
Conclusion
Our results show that the sEH can regulate mechanical hyperalgesia during inflammation by inactivating 8,9-EET, which sensitizes TRPA1-expressing nociceptors. Therefore we suggest that influencing the CYP450 pathway, which is actually highly considered to treat cardiovascular diseases, may cause pain side effects.

High attrition rates of novel anti-cancer drugs highlight the need for improved models to predict toxicity. Although polo-like kinase 1 (Plk1) inhibitors are attractive candidates for drug development, the role of Plk1 in primary cells remains widely unexplored. Therefore, we evaluated the utility of an RNA interference-based model to assess responses to an inducible knockdown (iKD) of Plk1 in adult mice. Here we show that Plk1 silencing can be achieved in several organs, although adverse events are rare. We compared responses in Plk1-iKD mice with those in primary cells kept under controlled culture conditions. In contrast to the addiction of many cancer cell lines to the non-oncogene Plk1, the primary cells' proliferation, spindle assembly and apoptosis exhibit only a low dependency on Plk1. Responses to Plk1-depletion, both in cultured primary cells and in our iKD-mouse model, correspond well and thus provide the basis for using validated iKD mice in predicting responses to therapeutic interventions.
Host cell invasion by the facultative intracellular pathogen Listeria monocytogenes requires the invasion protein InlB in many cell types. InlB consists of an N-terminal internalin domain that binds the host cell receptor tyrosine kinase Met and C-terminal GW domains that bind to glycosaminoglycans (GAGs). Met binding and activation is required for host cell invasion, while the interaction between GW domains and GAGs enhances this effect. Soluble InlB elicits the same cellular phenotypes as the natural Met ligand hepatocyte growth factor/scatter factor (HGF/SF), e.g. cell scatter. So far, little is known about the central part of InlB, the B-repeat. Here we present a structural and functional characterization of the InlB B-repeat. The crystal structure reveals a variation of the β-grasp fold that is most similar to small ubiquitin-like modifiers (SUMOs). However, structural similarity also suggests a potential evolutionary relation to bacterial mucin-binding proteins. The B-repeat defines the prototype structure of a hitherto uncharacterized domain present in over a thousand bacterial proteins. Generally, this domain probably acts as a spacer or a receptor-binding domain in extracellular multi-domain proteins. In cellular assays the B-repeat acts synergistically with the internalin domain conferring to it the ability to stimulate cell motility. Thus, the B-repeat probably binds a further host cell receptor and thereby enhances signaling downstream of Met.
Die Übertragung der semantischen Werte der deutschen Modalverben ins Portugiesische ist unstreitig ein komplexes Thema, mit dem sich alle, die mit Sprachen arbeiten, auseinandersetzen müssen: Übersetzer und Lexikographen sowie die am Übersetzungsunterricht und am Lehr- und Lernprozess von Deutsch und Portugiesisch als Fremdsprachen Beteiligten. Diese Komplexität ergibt sich weitgehend aus der multifunktionalen Natur dieser Verben, die das Ergebnis eines Entwicklungsprozesses ist, in dem den ursprünglich nicht-epistemischen Verwendungen eine weitere epistemische Lesart hinzugefügt wurde. Durch diese zusätzliche Bedeutungsdimension wird der Vertrauensgrad des Sprechers bezüglich des Äußerungsinhalts wiedergegeben. Das Verb sollen und die vielfältigen von ihm ausgelösten Interpretationen sind hierfür ein besonders anschauliches Beispiel. Für die Aktivierung einer bestimmten Lesart trägt eine Vielzahl kontextueller Elemente bei, die nicht ignoriert werden kann. Die Übersetzung von sollen erfordert daher eine Aufmerksamkeit auf diese Vielzahl von Faktoren, die – öfter als man es sich vorstellen kann – der Wahrnehmung von Übersetzern entgeht. Unser Beitrag, der auf einem Korpus von zeitgenössischen deutschen literarischen und pragmatischen Texten sowie deren portugiesischen Übersetzungen basiert, versucht dieser Komplexität Rechnung zu tragen.
PURPOSE: This study investigates the distortion of geometry of catheters and anatomy in acquired U/S images, caused by utilizing various stand-off materials for covering a transrectal bi-planar ultrasound probe in HDR and LDR prostate brachytherapy, biopsy and other interventional procedures. Furthermore, an evaluation of currently established water-bath based quality assurance (QA) procedures is presented.
MATERIAL AND METHODS: Image acquisitions of an ultrasound QA setup were carried out at 5 MHz and 7 MHz. The U/S probe was covered by EA 4015 Silicone Standoff kit, or UA0059 Endocavity balloon filled either with water or one of the following: 40 ml of Endosgel(®), Instillagel(®), Ultraschall gel or Space OAR™ gel. The differences between images were recorded. Consequently, the dosimetric impact of the observed image distortion was investigated, using a tissue equivalent ultrasound prostate phantom - Model number 053 (CIRS Inc., Norfolk, VA, USA).
RESULTS: By using the EA 4015 Silicone Standoff kit in normal water with sound speed of 1525 m/s, a 3 mm needle shift was observed. The expansion of objects appeared in radial direction. The shift deforms also the PTV (prostate in our case) and other organs at risk (OARs) in the same way leading to overestimation of volume and underestimation of the dose. On the other hand, Instillagel(®) and Space OAR™ "shrinks" objects in an ultrasound image for 0.65 mm and 0.40 mm, respectively.
CONCLUSIONS: The use of EA 4015 Silicone Standoff kit for image acquisition, leads to erroneous contouring of PTV and OARs and reconstruction and placement of catheters, which results to incorrect dose calculation during prostate brachytherapy. Moreover, the reliability of QA procedures lies mostly in the right temperature of the water used for accurate simulation of real conditions of transrectal ultrasound imaging.
There are many tools available that are used to evaluate a radiotherapy treatment plan, such as isodose distribution charts, dose volume histograms (DVH), maximum, minimum and mean doses of the dose distributions as well as DVH point dose constraints. All the already mentioned evaluation tools are dosimetric only without taking into account the radiobiological characteristics of tumors or OARs. It has been demonstrated that although competing treatment plans might have similar mean, maximum or minimum doses they may have significantly different clinical outcomes (Mavroidis et al. 2001). For performing a more complete treatment plan evaluation and comparison the complication-free tumor control probability (P+) and the biologically effective uniform dose (D ) have been proposed (Källman et al. 1992a, Mavroidis et al. 2000). The D concept denotes that any two dose distributions within a target or OAR are equivalent if they produce the same probability for tumor control or normal tissue complication, respectively (Mavroidis et al. 2001)...
Bei der chronisch venösen Insuffizienz (CVI) handelt es sich um einen im Bereich der unteren Extremität lokalisierten varikösen Symptomenkomplex bestehend aus Beschwerden wie Schmerzen, Schwere-, Spannungsgefühl, Juckreiz uvm. Dazu kommen stadienabhängig trophische Hautveränderungen, bedingt durch veränderte Kapillarmorphologie und -dichte. Diese Veränderungen können, je nach Ausprägungsgrad, von Hyperpigmentation über Dermatitis, Corona phlebectatica paraplantaris, Atrophie blanche bis hin zum floriden Ulcus cruris venosum reichen.
In der Bundesrepublik Deutschla nd leiden ca. 10-15 Millionen Menschen an einer manifesten CVI. Der CVI kommt aufgrund ihrer hohen Prävalenz eine hohe sozialmedizinische und sozialökonomische Bedeutung zu. Der pathophysiologische Mechanismus, der der CVI zugrunde liegt, ist bei Hinzukommen von begünstigenden Faktoren, z.B. Orthostasebelastung, in der Entwicklung von insuffizienten Venenabschnitten oder Insuffizienzpunkten im Bereich der Venenklappen oder anderen am Rücktransport des Blutes zum Herzen beteiligten Mechanismen zu suchen.
Eine Vielzahl von Therapieverfahren, wie etwa operative Eingriffe und Sklerosierungen, sind auf die Ausschaltung dieser Insuffizienzpunkte ausgerichtet. Diese Verfahren sind gründlich erforscht und durch klinische Studien wohldokumentiert. Ihre Grenzen liegen zum einen in möglichen Nebenwirkungen, zum anderen in der grundlegend chronisch degenerativen Natur der Erkrankung, deren Ursachen durch solche Therapieformen nicht erfasst werden.
Andere, nicht invasive und meist physikalische Therapieformen zielen auf eine Verbesserung der subjektiv empfundenen Lebensqualität ab. So ist etwa die Wirksamkeit von kalten hydrotherapeutischen Anwendungen wie Knie- oder Beingüssen, Wassertreten, Lehm-Wadenwickeln oder wechselwarmen Anwendungen nach Kneipp auf die subjektiven Beschwerden mehrfach beschrieben und in evidenten Studien belegt worden.
Wie es sich in diesem Zusammenhang mit der Wirksamkeit kalter Lehmpackungen, so wie sie als ortsgebundenes Heilmittel in Kurbädern Anwendung finden, verhält, ist hingegen bis dato nicht systematisch ergründet worden. Um diesen Mangel zu beheben, wurde die vorliegende randomisierte, kontrollierte Studie zur Wirksamkeit von kalten Lehmpackungen auf die Beine von CVI-Patienten entworfen. In dieser Studie wurden zum einen die Wirkungen dieser Behandlungsform auf die subjektiv erfassten Größen Lebensqualität, Schmerzen und Stauungsbeschwerden ermittelt, unter Anwendung des SF (short form) 36-Fragebogens und der visuellen Analogskala (VAS). Als Hauptzielgröße wurde die subjektiv empfundene Lebensqualität gewählt. Zum anderen wurden die objektiven Messparameter Knöchel-, Wadenumfang, transkutan gemessener Sauerstoffpartialdruck und die venöse Wiederauffüllzeit als Nebenzielgrößen erfasst.
Im direkten Anschluss an die Therapie lassen sich signifikante Verbesserungen der subjektiven Messparameter verzeichnen. Diese fallen bei den krankheitsspezifischen Faktoren und im körperlichen Lebensqualitätsprofil deutlicher aus als im psychischen Lebensqualitätsprofil. Innerhalb des körperlichen Lebensqualitätsprofils wiederum zeigen sich die deutlichsten Verbesserungen bei der körperlichen Rollenfunktion, gefolgt von den körperlichen Schmerzen. Das psychische Lebensqualitätsprofil weist die deutlichste Verbesserung bei der emotionalen Rollenfunktion auf. Diese Ergebnisse lassen sich in der vorliegenden Kombination vor dem Hintergrund der Beobachtungen erklären, dass zum einen die CVI eine chronisch degenerative körperliche Erkrankung ist, deren Effekte sich vornehmlich in körperlichen und psychischen Funktionalitätseinbußen manifestieren, und dass sich zum anderen das Patientenkollektiv der Studie durch fortgeschrittenes Alter und einen insgesamt unterdurchschnittlichen gesundheitlichen Allgemeinzustand auszeichnet. Alter und gesundheitlicher Allgemeinzustand wiederum gehen in erster Linie mit dauerhaft empfundenen Beeinträchtigungen der körperlichen und psychischen Rollenfunktionen einher. Durch die Lehmpackungen gelingt nun eine Reduktion der Schmerzen und anderer Beeinträchtigungen der körperlichen Befindlichkeit (Schweregefühl, Juckreiz). Diese Verbesserungen machen es dem Patienten möglich, seine an ihn gestellten Rollenerwartungen besser zu erfüllen.
Die erreichten positiven Effekte auf die subjektiven Parameter tendieren nach drei Monaten wieder in Richtung Ausgangswert, ohne diesen jedoch zu erreichen, so dass sich eine gewisse Nachhaltigkeit des Therapieeffektes zeigt. Die objektiven Messparameter tendieren in Richtung einer Verbesserung (Ausnahme: venöse Wiederauffüllzeit rechts), erreichen jedoch kein hinreichendes Signifikanzniveau. Um diesbezüglich ein aussagekräftiges Ergebnis zu erhalten, wären ggf. eine Vergrößerung der Stichprobe und andere bzw. verfeinerte Messmethoden angebracht.
Using an electrophysiological assay the activity of NhaA was tested in a wide pH range from pH 5.0 to 9.5. Forward and reverse transport directions were investigated at zero membrane potential using preparations with inside-out and right side-out-oriented transporters with Na+ or H+ gradients as the driving force. Under symmetrical pH conditions with a Na+ gradient for activation, both the wt and the pH-shifted G338S variant exhibit highly symmetrical transport activity with bell-shaped pH dependences, but the optimal pH was shifted 1.8 pH units to the acidic range in the variant. In both strains the pH dependence was associated with a systematic increase of the Km for Na+ at acidic pH. Under symmetrical Na+ concentration with a pH gradient for NhaA activation, an unexpected novel characteristic of the antiporter was revealed; rather than being down-regulated, it remained active even at pH as low as 5. These data allowed a transport mechanism to advance based on competing Na+ and H+ binding to a common transport site and a kinetic model to develop quantitatively explaining the experimental results. In support of these results, both alkaline pH and Na+ induced the conformational change of NhaA associated with NhaA cation translocation as demonstrated here by trypsin digestion. Furthermore, Na+ translocation was found to be associated with the displacement of a negative charge. In conclusion, the electrophysiological assay allows the revelation of the mechanism of NhaA antiport and sheds new light on the concept of NhaA pH regulation.
Explosive volcanism affects weather and climate. Primary volcanic ash particles which act as ice nuclei (IN) can modify the phase and properties of cold tropospheric clouds. During the Eyjafjallajökull volcanic eruption we have measured the highest ice nucleus number concentrations (>600 L) in our record of 2 years of daily IN measurements in central Germany. Even in Israel, located about 5000 km away from Iceland, IN were as high as otherwise only during desert dust storms. These measurements are the only ones available on the properties of IN in the Eyjafjallajökull plume. The measured high concentrations and high activation temperature (−8 °C) point to an important impact of volcanic ash on microphysical and radiative properties of clouds through enhanced glaciation.
Single crystals suitable for X-ray diffraction of (tBu2P)3Ga (monoclinic, space group Cc) were obtained from GaCl3 and two equivalents of Li[PtBu2] at room temperature in benzene. The phosphanylgallane (tBu2P)3Ga was also produced via a one-pot approach by reaction of GaCl3 with three or more than three equivalents of Li[PtBu2]. However, treatment of one equivalent of GaCl3 with one equivalent of Li[PtBu2] and subsequent protolysis yielded [tBu2PH2][tBu2P(GaCl3)2 - Li(Cl3Ga)2PtBu2]. Single crystals of this phosphonium salt (monoclinic, space group Cc) were obtained from benzene at room temperature.
A new gymnophthalmid lizard of the genus Anadia Gray, 1845 is described from the summit of Abakapá-tepui, Bolívar State, Venezuela, between 2200-2242 m elevation. The new species, Anadia mcdiarmidi sp. nov., is endemic to the Chimantá Massif and seemingly also occurs on Amurí-tepui and Murei-tepui. The new taxon is mainly distinguished from all known congeners by the following combination of characters: body fairly robust, dorsal scales small and quadrangular, middorsal scales 53-57, suboculars large, subequal in size, with sometimes one scale slightly protruding downward between 4th and 5th supralabial, nasal entire, without sub-nostril groove, body uniform beige or greyish to bluish brown in life, devoid of any conspicuous pattern in males, venter immaculate golden grey in life, femoral pores 9-10 on each side in males, preanal pores absent, hemipenis globose, weakly bilobed, bordered by numerous fl ounces (>20) bearing comblike rows of minute weakly mineralized spinules. The presence of a species of Anadia, a primarily Andean genus, on the top of tepuis is of considerable interest to the understanding of the Pantepui biogeography.
To examine their luminescence behavior, two air-stable BN addition compounds were synthesized by the reaction of 5-fluoro-2-(2′-pyridyl)indole with 1,4- and 1,3-bis(bromo(methyl)boryl)benzene, respectively. Both BN adducts are luminescent. Their emission maxima (1,3-substituted BN adduct: 495 nm; 1,4-substituted BN adduct: 497 nm) are comparable with the value (490 nm) of the related mono-borylated benzene species, which is composed of a BPh2 fragment and a 5-fluoro-2-(2′-pyridyl) indole unit. The starting materials 1,4- and 1,3-bis(bromo(methyl)boryl)benzene were accessible by treatment of 1,4- or 1,3-bis(dibromoboryl)benzene with two equivalents of SnMe4. In addition, the results of the X-ray structure analyses of the B,B′-bis-5-fluoro-2-(2′-pyridyl)indolyl-complexed meta-bismethylborylbenzene fragment (9, triclinic, P1̅) as well as of 5-chloro-2-(2′-pyridyl)indole (2, monoclinic, P21/c) and 5-fluoro-2-(2′-pyridyl)indole (1, orthorhombic, Pca21) are reported. The pyridylindole derivatives of this approach were synthesized by an optimized two-step procedure from 2-acetylpyridine and 4-fluoro- or 4-chlorophenylhydrazine hydrochloride.
Einleitung:
Wahrscheinlich hat es seit den 1970er Jahren nicht mehr so viele kritische Initiativen von Geographiestudierenden an Hochschulen in deutschsprachigen Ländern gegeben wie heute. Ihre Aktivitäten reichen von Lesekreisen, Tutorien, Exkursionen, Film- und Vortragsreihen bis zu politischen Aktionen, wobei viele Initiativen über den Arbeitskreis (AK) Kritische Geographie vernetzt sind. Parallel zu dieser erfreulichen Beobachtung findet ein fundamentaler Umbau der Hochschule statt. Die zunehmende Ökonomisierung des Studiums durch den Bologna-Prozess birgt die Gefahr, Gesellschaftskritik auf dem Altar der Verwertbarkeit und der "Praxisrelevanz" zu opfern. Diese Entwicklungen geben Anlass, das Verhältnis zwischen der Arbeit kritischer Initiativen und den reformierten Universitäten kritisch zu hinterfragen und damit den Zusammenhang zwischen Bologna-Prozess, Neoliberalisierung und kritischer Wissenschaft für die geographische Hochschullehre aus einer studentischen Perspektive zu beleuchten, wird doch ein nicht unbedeutender Teil der kritischen Geographie von Studierenden getragen.
Im Folgenden werden wir kurz auf den Bologna-Prozess eingehen, anschließend die Fragestellung und das methodische Vorgehen erläutern, um darauf aufbauend die empirischen Ergebnisse vorzustellen und zu diskutieren.
The vespid fauna of Greater Puerto Rico is reviewed (Hymenoptera: Vespidae). Polistes crinitus americanus (Fabricius, 1775) and P. crinitus multicolor (Olivier, 1792) are both reduced to synonyms of nominotypical P. crinitus (Felton, 1765), revised status; Zeta abdominale hispaniolae (Bequaert and Salt, 1931) and Zeta abdominale ornatum (de Saussure, 1855) are both reduced to synonyms of nominotypical Zeta abdominale (Drury, 1770), revised status; and Zethus rufinodus monensis Bohart and Stange, 1965, and Zethus rufinodus virginicus Bohart and Stange, 1965, are both reduced to synonyms of nominotypical

from Puerto Rico. The presence of Pachodynerus guadulpensis (de Saussure, 1853) in Puerto Rico is confirmed. An analysis of the composition of the Puerto Rican vespid fauna is presented.
Die Internationalisierung der deutschen Hochschulen nahm in den letzten Jahren stark zu. Umgang mit Studierenden aus unterschiedlichen Kulturen bedeutet für Lehrende längst Alltag. Nicht immer jedoch verläuft die Kommunikation zwischen Angehörigen unterschiedlicher Kulturen reibungslos. Um möglichen Schwierigkeiten entgegenzuwirken, setzen einige Universitäten interkulturelle Trainings ein zur Sensibilisierung für interkulturelle Unterschiede. Die Autoren haben im Rahmen eines hochschuldidaktischen Weiterbildungsprogramms für Lehrende ein interkulturelles Training entwickelt und eingesetzt. Über den Aufbau und die Ziele des Trainings wird im vorliegenden Artikel berichtet. Weiterhin wird ein Untersuchungsdesign vorgestellt, mit welchem der Einfluss von Kultur auf die Online-Kommunikation in der Lehre untersucht wurde.
Acupuncture is a therapy based on sensory stimulation of the human
body by means of metal needles. The exact underlying mechanisms of
acupuncture have not been clarified so far. Functional magnetic
resonance imaging (fMRI) has become an important tool in
acupuncture research. Standard acupuncture needles, which are made
of ferromagnetic steel, however, are problematic in
acupuncture-fMRI studies for several reasons, such as attraction
by the scanner's magnetic field, significant image distortions and
signal-dropouts, when positioned close to the head or even heating
due to absorption of radio frequency (RF). The aim of this study
was to compare two novel types of acupuncture needles with a
standard needle for their effect on MRI image quality. The
standard needle severely reduced image quality, when located
inside the RF coil. The nonferromagnetic metal needle may pose a
risk due to RF heating, while the plastic needle has a
significantly larger diameter. In conclusion, our recommendations
are: (1) standard needles should not be used in MRI; (2)
Nonferromagnetic metal needles seem to be the best choice for
acupoints outside of the transmitter coil; and (3) only plastic
needles are suited for points inside the coil. Laser acupuncture
may be a safe alternative, too.
Ziel der vorliegenden Arbeit war es die Einflussmöglichkeiten von Umweltverbänden im energiepolitischen Entscheidungsprozess in Deutschland und in Südkorea zu vergleichen. Diese Studie untersuchte exemplarisch den BUND und die KFEM hinsichtlich ihrer Einflusschancen auf die Atompolitik in beiden Staaten innerhalb eines bestimmten Zeitraumes (vom Tschernobyl-Reaktorunfall 1986 bis 2007). Ein besonderer Fokus galt dabei den Faktoren, die auf unterschiedlichen Ebenen diese Einflusschancen herstellen bzw. minimieren...
This paper compares the shareholder-value-maximizing capital structure and pricing policy of insurance groups against that of stand-alone insurers. Groups can utilise intra-group risk diversification by means of capital and risk transfer instruments. We show that using these instruments enables the group to offer insurance with less default risk and at lower premiums than is optimal for standalone insurers. We also take into account that shareholders of groups could find it more difficult to prevent inefficient overinvestment or cross-subsidisation, which we model by higher dead-weight costs of carrying capital. The tradeoff between risk diversification on the one hand and higher dead-weight costs on the other can result in group building being beneficial for shareholders but detrimental for policyholders.

In modern regulation regimes, such as the future standard Solvency II in the EU, insurance pricing is liberalized and risk-based capital requirements will be introduced. In many economies in Asia and Latin America, on the other hand, supervisors require the prior approval of policy conditions and insurance premiums, but do not conduct risk-based capital regulation. This paper compares the outcome of insurance rate regulation and risk-based capital requirements by deriving stock insurers’ best responses. It turns out that binding price floors affect insurers’ optimal capital structures and induce them to choose higher safety levels. Risk-based capital requirements are a more efficient instrument of solvency regulation and allow for lower insurance premiums, but may come at the cost of investment efforts into adequate risk monitoring systems. The paper derives threshold values for regulator’s investments into risk-based capital regulation and provides starting points for designing a welfare-enhancing insurance regulation scheme.

Background: Even in the presence of oxygen, malignant cells often highly depend on glycolysis for energy generation, a phenomenon known as the Warburg effect. One strategy targeting this metabolic phenotype is glucose restriction by administration of a high-fat, low-carbohydrate (ketogenic) diet. Under these conditions, ketone bodies are generated serving as an important energy source at least for non-transformed cells. Methods: To investigate whether a ketogenic diet might selectively impair energy metabolism in tumor cells, we characterized in vitro effects of the principle ketone body 3-hydroxybutyrate in rat hippocampal neurons and five glioma cell lines. In vivo, a non-calorie-restricted ketogenic diet was examined in an orthotopic xenograft glioma mouse model. Results: The ketone body metabolizing enzymes 3-hydroxybutyrate dehydrogenase 1 and 2 (BDH1 and 2), 3-oxoacid-CoA transferase 1 (OXCT1) and acetyl-CoA acetyltransferase 1 (ACAT1) were expressed at the mRNA and protein level in all glioma cell lines. However, no activation of the hypoxia-inducible factor-1alpha (HIF-1alpha) pathway was observed in glioma cells, consistent with the absence of substantial 3-hydroxybutyrate metabolism and subsequent accumulation of succinate. Further, 3-hydroxybutyrate rescued hippocampal neurons from glucose withdrawal-induced cell death but did not protect glioma cell lines. In hypoxia, mRNA expression of OXCT1, ACAT1, BDH1 and 2 was downregulated. In vivo, the ketogenic diet led to a robust increase of blood 3-hydroxybutyrate, but did not alter blood glucose levels or improve survival. Conclusion: In summary, glioma cells are incapable of compensating for glucose restriction by metabolizing ketone bodies in vitro, suggesting a potential disadvantage of tumor cells compared to normal cells under a carbohydrate-restricted ketogenic diet. Further investigations are necessary to identify co-treatment modalities, e.g. glycolysis inhibitors or antiangiogenic agents that efficiently target non-oxidative pathways.
Seroconversion rates following influenza vaccination in patients with hematologic malignancies after hematopoietic stem cell transplantation (HSCT) are known to be lower compared to healthy adults. The aim of our diagnostic study was to determine the rate of seroconversion after 1 or 2 doses of a novel split virion, inactivated, AS03-adjuvanted pandemic H1N1 influenza vaccine (A/California/7/2009) in HSCT recipients (ClinicalTrials.gov Identifier: NCT01017172). Blood samples were taken before and 21 days after a first dose and 21 days after a second dose of the vaccine. Antibody (AB) titers were determined by hemagglutination inhibition assay. Seroconversion was defined by either an AB titer of ≤1:10 before and ≥1:40 after or ≥1:10 before and ≥4-fold increase in AB titer 21 days after vaccination. Seventeen patients (14 allogeneic, 3 autologous HSCT) received 1 dose and 11 of these patients 2 doses of the vaccine. The rate of seroconversion was 41.2% (95% confidence interval [CI] 18.4-67.1) after the first and 81.8% (95% CI 48.2-97.7) after the second dose. Patients who failed to seroconvert after 1 dose of the vaccine were more likely to receive any immunosuppressive agent (P = .003), but time elapsed after or type of HSCT, age, sex, or chronic graft-versus-host disease was not different when compared to patients with seroconversion. In patients with hematologic malignancies after HSCT the rate of seroconversion after a first dose of an adjuvanted H1N1 influenza A vaccine was poor, but increased after a second dose.
Background: Although being considered as a rarely observed HIV-1 protease mutation in clinical isolates, the L76V-prevalence increased 1998-2008 in some European countries most likely due to the approval of Lopinavir, Amprenavir and Darunavir which can select L76V. Beside an enhancement of resistance, L76V is also discussed to confer hypersusceptibility to the drugs Atazanavir and Saquinavir which might enable new treatment strategies by trying to take advantage of particular mutations. Results: Based on a cohort of 47 L76V-positive patients, we examined if there might exist a clinical advantage for L76V-positive patients concerning long-term success of PI-containing regimens in patients with limited therapy options. Genotypic- and phenotypic HIV-resistance tests from 47 mostly multi-resistant, L76V-positive patients throughout Germany were accomplished retrospectively 1999-2009. Five genotype-based drug-susceptibility predictions received from online interpretation-tools for Atazanavir, Saquinavir, Amprenavir and Lopinavir, were compared to phenotype-based predictions that were determined by using a recombinant virus assay along with a Virtual Phenotype™(Virco). The clinical outcome of the L76V-adapted follow-up therapy was determined by monitoring viral load for 96 weeks. Conclusions: In this analysis, the mostly used interpretation systems overestimated the L76V-mutation concerning Atazanavir- and SQV resistance. In fact, a clear benefit in drug susceptibility for these drugs was observed in phenotype analysis after establishment of L76V. More importantly, long-term therapy success was significantly higher in patients receiving Atazanavir and/or Saquinavir plus one L76V-selecting drug compared to patients without L76V-selecting agents (p = 0.002). In case of L76V-occurrence ATV and/or SQV may represent encouraging options for patients in deep salvage situations.

Der Unterzeichnende – oft genug hat er mit scharfer Rezensentenklinge gefochten – darf eine Kollegin rühmen, die nach ihrer Pensionierung als Forschungsdirektorin am C.N.R.S. 2011 in Bourges mit einem internationalen Kongress geehrt wurde (Église et État, Église ou État? Les clercs et la genèse de l’État moderne) und die in den letzten Jahren als Summa ihrer Forschungen zum großen abendländischen Schisma gleich drei Bände vorlegen konnte, von denen zwei wiederum Konzilien jener Zeit mit Jubiläumsdatum galten: So gab Hélène Millet 2009 die Akten einer Tagung heraus, die, wesentlich von ihr konzipiert und organisiert, im Jahr zuvor aus Anlass des sechsten Centenariums des Konzils von Perpignan an ebendiesem Ort stattgefunden hatte 1 . Damit rückte sie jene Synode Benedikts XIII. gegen das anstehende und ihn bedrohende Pisanum in klareres Forschungslicht, die im Deutschen seit der Edition ihrer Akten durch Franz Ehrle (1889/1900) als "Afterconcil von Perpignan" allenfalls randhaft zur Kenntnis genommen wurde. H. Millet hat danach selbst noch wiederholt zu Inhalt und Ertrag dieses Kolloquiums Stellung genommen, so schriftlich in "cristianesimo nella storia" (29, 2008, S. 219–229) und mündlich im Rahmen einer Tagung über den "Languedoc médiéval" im Februar 2011 in Montpellier. 2009 war zudem das Jahr, in dem sie unter dem Titel "L’Église du Grand Schisme 1378–1417" eine Auswahl ihrer – meist frankreichzentrierten – Studien zur Kirchenspaltung vorlegte 2 . Immer wieder lassen sie ihre Kompetenz im prosopographisch-biographischen Bereich und damit auch ihre – computergestützten – Erfahrungen in der Kanoniker- und Kapitelforschung aufscheinen, aus denen wiederum das von ihr inspirierte und mittlerweile wohletablierte Unternehmen der Fasti Ecclesiae Gallicanae großen Nutzen gezogen hat und zieht. ...
Es ist schon erstaunlich: Da richtet ein Verlag eine sich ausschließlich an Fortgeschrittene wendende Reihe ein, vertreibt die über recht spezielle Themen handelnden Bände zu für Privatleute prohibitiven Preisen zwischen 100 und 160 Euro und kann dennoch innerhalb von nur vier Jahren (bis Sept. 2010) nicht weniger als 23 Publikationen vorlegen. Offensichtlich rechnet sich das bei einem international agierenden und mit den "Companions" wohl primär auf wissenschaftliche Bibliotheken zielenden Haus: eine gute Sache bei selbst für diese Institutionen schlechtem Preis. Ob es – bei einer bisherigen gewissen Schwerpunktsetzung in Spätmittelalter und Früher Neuzeit – um theologische Quodlibeta im 13. und 14. Jahrhundert, um Wyclif und Gerson oder um katholische Aufklärung geht, stets enthalten die Bände acht bis fünfzehn Beiträge einschlägig ausgewiesener Autoren samt Einführung, (teilweiser) Konklusion und Auswahlbibliographie der Herausgeber. Jedes Buch soll die Summe des Forschungsstands darstellen, und recht selbstbewusst spricht man im Untertitel der Reihe von "A series of handbooks and reference works". ...
8th International Conference on Nuclear Physics at Storage Rings Stori11, October 9-14, 2011 Laboratori Nazionale di Frascati, Italy.
Storage rings offer the possibility of measuring proton- and alpha-induced reactions in inverse kinematics. The combination of this approachwith a radioactive beamfacility allows, in principle, the determination of the respective cross sections for radioactive isotopes. Such data are highly desired for a better understanding of astrophysical nucleosynthesis processes like the p-process. A pioneering experiment has been performed at the Experimental Storage Ring (ESR) at GSI using a stable 96Ru beam at 9-11 AMeV and a hydrogen target. Monte-Carlo simulations of the experiment were made using the Geant4 code. In these simulations, the experimental setup is described in detail and all reaction channels can be investigated. Based on the Geant4 simulations, a prediction of the shape of different spectral components can be performed. A comparison of simulated predictions with the experimental results shows a good agreement and allows the extraction of the cross section.

Ca. 3 Millionen Erwachsene in der Bundesrepublik Deutschland leiden unter Tinnitus, wobei eine bei jedem dieser Patienten zur Heilung führende Therapie bisher noch nicht existiert. Ansatzpunkt einer neuartigen Therapie ist die Wiederherstellung des normalen elektrischen Entladungsmusters im Hörnerv mittels elektrischer Stimulation. Hiermit berichten wir über unsere ersten Erfahrungen mit dem Tinnelec, einem Implantat mit einer einzelnen Stimulations-Elektrode die in der Rundfensternische platziert wird.
Zurzeit haben wir 4 einseitig ertaubten Patienten mit Tinnitus auf dem betroffenen Ohr jeweils ein Tinnelec-System implantiert. Die Dauer des Tinnitus betrug mindestens ein Jahr und gängige Tinnitus-Therapien wie z.B. Infusionstherapie waren erfolglos geblieben. Ein psychogener Tinnitus wurde ausgeschlossen. Der durch den Tinnitus verursachte Leidensdruck wurde anhand einer VAS Scala (Visuelle Analog Scala) und eines Tinnitus-Handicap-Inventory (THI) Fragebogens beurteilt. Die Reizapplikation betrug mind. 4 Stunden täglich. Als Stimulationsparameter wurde eine Reizmusterannäherung an den Tinnitus angestrebt.
Bei drei Patienten wurde unter der Stimulation der Tinnitus erträglicher, eine zeitweise komplette Unterdrückung des Tinnitus schon innerhalb der ersten Therapie-Wochen wurde jedoch nur in einem der Fälle berichtet. Diese Ergebnisse wurden auch durch das THI und VAS unterstützt.
Die Tinnelec-Implantation erscheint für Tinnitus Erfolg versprechend zu sein. Weitere Studien bei Tinnitus-Patienten ohne zusätzliche Hörbeeinträchtigung sind jedoch notwendig bis endgültige Schlussfolgerungen betreffend dieses Implantats gezogen werden können. The cochlear implant (CI) represents, for almost 25 years now, the gold standard in the treatment of children born deaf and for postlingually deafened adults. These devices thus constitute the greatest success story in the field of ‘neurobionic’ prostheses. Their (now routine) fitting in adults, and especially in young children and even babies, places exacting demands on these implants, particularly with regard to the biocompatibility of a CI’s surface components. Furthermore, certain parts of the implant face considerable mechanical challenges, such as the need for the electrode array to be flexible and resistant to breakage, and for the implant casing to be able to withstand external forces. As these implants are in the immediate vicinity of the middle-ear mucosa and of the junction to the perilymph of the cochlea, the risk exists – at least in principle – that bacteria may spread along the electrode array into the cochlea. The wide-ranging requirements made of the CI in terms of biocompatibility and the electrode mechanism mean that there is still further scope – despite the fact that CIs are already technically highly sophisticated – for ongoing improvements to the properties of these implants and their constituent materials, thus enhancing the effectiveness of these devices. This paper will therefore discuss fundamental material aspects of CIs as well as the potential for their future development. Six p53 wild-type cancer cell lines from infrequently p53-mutated entities (neuroblastoma, rhabdomyosarcoma, and melanoma) were continuously exposed to increasing concentrations of the murine double minute 2 inhibitor nutlin-3, resulting in the emergence of nutlin-3-resistant, p53-mutated sublines displaying a multi-drug resistance phenotype. Only 2 out of 28 sublines adapted to various cytotoxic drugs harboured p53 mutations. Nutlin-3-adapted UKF-NB-3 cells (UKF-NB-3rNutlin10 μM, harbouring a G245C mutation) were also radiation resistant. Analysis of UKF-NB-3 and UKF-NB-3rNutlin10 μM cells by RNA interference experiments and lentiviral transduction of wild-type p53 into p53-mutated UKF-NB-3rNutlin10 μM cells revealed that the loss of p53 function contributes to the multi-drug resistance of UKF-NB-3rNutlin10 μM cells. Bioinformatics PANTHER pathway analysis based on microarray measurements of mRNA abundance indicated a substantial overlap in the signalling pathways differentially regulated between UKF-NB-3rNutlin10 μM and UKF-NB-3 and between UKF-NB-3 and its cisplatin-, doxorubicin-, or vincristine-resistant sublines. Repeated nutlin-3 adaptation of neuroblastoma cells resulted in sublines harbouring various p53 mutations with high frequency. A p53 wild-type single cell-derived UKF-NB-3 clone was adapted to nutlin-3 in independent experiments. Eight out of ten resulting sublines were p53-mutated harbouring six different p53 mutations. This indicates that nutlin-3 induces de novo p53 mutations not initially present in the original cell population. Therefore, nutlin-3-treated cancer patients should be carefully monitored for the emergence of p53-mutated, multi-drug-resistant cells.
Based on a collection of spiders obtained during ecological fieldwork in 2009 and an extensive literature review, we summarize the current state of knowledge of spider biodiversity on the island of Maio. The total number of species reported from Maio is now 46, representing 18 families and including 16 species (35%) endemic to the Cape Verde Islands. The family Dictynidae (meshweb spiders), represented by the saline-adapted Devade cf. indistincta, is reported for the first time from Cape Verde.
In dieser Arbeit wurden zwei Schlüsselenzyme des Energiestoffwechsels in Archaeen im Hinblick auf ihre funktionellen, spektroskopischen und strukturellen Eigenschaften untersucht. Die Heterodisulfid-Reduktase (Hdr) katalysiert die Reduktion des terminalen Elektronenakzeptors CoM-S-S-CoB zu CoM-SH (Coenzym M) und CoB-SH (Coenzym B) und spielt eine Schlüsselrolle im zentralen Energie-konservierenden Prozess von methanogenen Archaeen. Hdr existiert in Form von zwei unterschiedlichen Enzymen: HdrDE und HdrABC. Beide weisen ein charakteristisches Cystein-reiches Sequenzmotiv (CCG-Domäne) auf, welches als Bindestelle für ein ungewöhliches [4Fe-4S]-Zentrum dient. Frühere Studien zeigten, dass das [4Fe-4S]-Zentrum in der Untereinheit HdrB lokalisiert ist und als zentraler Bestandteil des aktiven Zentrums die Fähigkeit besitzt, ein Thiyl-Radikal zu binden. Darauf aufbauend wurden genetische, spektroskopische und strukturelle Untersuchungen überwiegend am H2:Heterodisulfid-Oxidoreduktase-Komplex (Mvh:Hdr) aus Methanothermobacter marburgensis oder an der heterolog produzierten Untereinheit HdrB durchgeführt. Das Reinigungsprotokoll des Mvh:Hdr-Komplexes wurde für Kristallisationsexperimente und für ENDOR- und Mössbauer-spektroskopische Studien optimiert. Eine Kristallisation des Mvh:Hdr-Komplexes gelang nicht; doch konnten Kristalle der Heterodisulfid-Reduktase-assoziierten Hydrogenase (Mvh) bis zu einer Auflösung von 3.34 Å vermessen und mit Hilfe der anomalen Information der Elektronentransferweg zwischen den [Fe-S]-Clustern definiert werden. Ergänzende elektronenmikroskopische Studien zeigten einen unsymmetrischen Aufbau des Komplexes. DesWeiteren wurde die Untereinheit HdrB aus M. marburgensis in Methanosarcina acetivorans heterolog produziert und seine Funktionalität kinetisch und spektroskopisch nachgewiesen. Ferner wurde HdrB in Escherichia coli heterolog produziert und gereinigt, um Kristallisationsexperimente durchzuführen und es für ENDOR- und Mössbauer-Studien verfügbar zu machen. Um HdrB spektroskopisch zu vergleichen, wurde eine Untereinheit der Succinat:Chinon Oxidoreduktase (SdhE) aus Sulfolobus solfataricus ebenfalls heterolog in E. coli produziert und mittels ENDOR-Spektroskopie charakterisiert. Ein grundlegender Prozess des biogeochemischen Schwefelkreislaufes ist die dissimilatorische Sulfat-Reduktion, in der Sulfat (SO4 2􀀀) zu Schwefelwasserstoff (H2S) umgewandelt wird. Die dissimilatorische Sulfit-Reduktase (dSir), das Schlüsselenzym im Energiestoffwechsel der Sulfat-Reduzierer, besitzt einen einzigartigen Sirohäm-[4Fe-4S]-Cofaktor, der die Reduktion von Sulfit (SO3 2􀀀) zu H2S in einem 6-Elektronen-Schritt katalysiert. Um diesen Mechanismus zu untersuchen, wurden kinetische, spektroskopische und röntxi Zusammenfassung genkristallographische Methoden angewandt. Die Kristallstrukturen von dSir aus Archaeoglobus fulgidus wurden im Komplex mit Sulfit, Sulfid (S2􀀀), Kohlenmonoxid (CO), Cyanid (CN􀀀), Nitrit (NO2􀀀), Nitrat (NO3 􀀀) und Phosphat (PO4 3􀀀) gelöst. Aktivitätstest und analytische Studien zeigten, dass dSir von A. fulgidus neben Sulfit und Nitrit auch Thiosulfat und Trithionat reduziert und Letztere auch als Intermediate entstehen. Auf dieser Basis wurde ein 3-Stufen-Mechanismus postuliert, wobei jede Stufe aus einem 2-Elektronentransfer, einer Aufnahme von zwei Protonen und einer Dehydrationsreaktion besteht. Im Vergleich zur assimilatorischen Sulfit-Reduktase (aSir) aus E. coli zeigt die dSir-Struktur einen veränderten Substratkanal, eine Rotation des Sulfits um 60° und beträchtliche Konformationsänderungen der katalytischen Reste Arga170 und Lysa211. Aufgrund dieser Änderungen kann ausschließlich in dSir ein weiteres Sulfit-Molekül in van-der-Waals-Kontakt zum an das Sirohäm-gebundene Sulfit oder Schwefel-Sauerstoff-Zwischenprodukt platziert werden, das nötig ist, um Thiosulfat und Trithionat zu synthetisieren.
IL-22 is an immunoregulatory cytokine displaying pathological functions in models of autoimmunity like experimental psoriasis. Understanding molecular mechanisms driving IL-22, together with knowledge on the capacity of current immunosuppressive drugs to target this process, may open an avenue to novel therapeutic options. Here, we sought to characterize regulation of human IL22 gene expression with focus on the established model of Jurkat T cells. Moreover, effects of the prototypic immunosuppressant cyclosporin A (CsA) were investigated. We report that IL-22 induction by TPA/A23187 (T/A) or αCD3 is inhibited by CsA or related FK506. Similar data were obtained with peripheral blood mononuclear cells or purified CD3(+) T cells. IL22 promoter analysis (-1074 to +156 bp) revealed a role of an NF-AT (-95/-91 nt) and a CREB (-194/-190 nt) binding site for gene induction. Indeed, binding of CREB and NF-ATc2, but not c-Rel, under the influence of T/A to those elements could be proven by ChIP. Because CsA has the capability to impair IκB kinase (IKK) complex activation, the IKKα/β inhibitor IKKVII was evaluated. IKKVII likewise reduced IL-22 induction in Jurkat cells and peripheral blood mononuclear cells. Interestingly, transfection of Jurkat cells with siRNA directed against IKKα impaired IL22 gene expression. Data presented suggest that NF-AT, CREB, and IKKα contribute to rapid IL22 gene induction. In particular the crucial role of NF-AT detected herein may form the basis of direct action of CsA on IL-22 expression by T cells, which may contribute to therapeutic efficacy of the drug in autoimmunity.
Ziel der vorliegenden tierexperimentellen Studie am narkotisierten Hausschwein
war die Untersuchung der Effekte von hyperoxischer Beatmung (Beatmung mit
reinem Sauerstoff; FiO2 1,0; HV) auf die Überlebenszeit bei schwerer
Methämoglobinämie. Hiermit sollten die Effekte der hyperoxischen Beatmung
bei einer Einschränkung des Sauerstoffangebots ohne gleichzeitige Reduktion
des Hämatokrit-Wertes, und damit der Blutviskosität untersucht werden.
Zielparameter waren die Überlebenszeit und die Überlebensrate während eines
6-stündigen Beobachtungszeitraums, sowie Parameter der Makrohämodynamik,
des O2-Transportes und der globalen und lokalen Gewebeoxygenierung.
Bei 14 gesunden Hausschweinen in Allgemeinanästhesie wurde während
Beatmung mit Raumluft eine Methämoglobinämie (60% des Gesamt-Hb)
induziert und aufrechterhalten. Anschließend wurden die Tiere in zwei
Studiengruppen randomisiert: 7 Tiere wurden weiterhin mit Raumluft (FiO2 0,21)
beatmet, 7 Tiere hyperoxisch, d.h. mit reinem O2 (FiO2 1,0).
In der Folge wurden die Tiere für maximal 6 Stunden ohne weitere Intervention
beobachtet.
Die 60% Methämoglobinämie führte bei allen Tieren zu einer deutlichen
Verschlechterung von O2-Transportes, zu manifester Gewebehypoxie und – bei
Fortführung der Beatmung mit Raumluft (FiO2 0.21) – zum Tod der Tiere
innerhalb kurzer Zeit (max. 2h 40 min). Durch Beatmung mit reinem Sauerstoff
gelang es zwar die Überlebenszeit gegenüber den mit Raumluft beatmeten
Tieren signifikant um bis zu 2 h zu verlängern. Allerdings blieb die 6 hÜberlebensrate
unbeeinflusst: Keines der 14 Tiere überlebte den 6 h-
Beobachtungszeitraum.
Die Effekte der hyperoxischen Beatmung (FiO2 1.0) während
Methämoglobinämie (d.h. Hypoxämie bei normalem Hämatokrit) waren somit
deutlich geringer ausgeprägt als die in früheren Untersuchungen unserer
Arbeitsgruppe beschriebenen Effekte währen normovolämischer Anämie (d.h.
Hypoxämie bei verringertem Hämatokrit). Während der physikalisch gelöste O2
bei anämischer Hypoxie zu einer biologisch exzellent verfügbaren O2-
Ressource avanciert, die Gewebeoxygenierung nachweislich verbessert, und
somit Überlebenszeit als auch –rate signifikant erhöht, ist dieser Effekt
während hypoxischer Hypoxie nicht zu beobachten.
Diese Abhängigkeit der gewünschten positiven Effekte einer Hyperoxie vom
jeweils vorherrschenden Hämatokrit ist erklärbar durch die hyperoxische
Vasokonstriktion, welche während Methämoglobinämie zu einer zusätzlichen
Verschlechterung der Mikrozirkulation, und einer weiteren Reduktion von
regionalem O2- Angebot und Gewebeoxygenierung führt. Die Beatmung mit
reinem O2 führt zu einer generalisierten arteriolären Vasokonstriktion, die durch
Arachidonsäuremetabolite und eine reduzierte endotheliale NO-Freisetzung
bedingt ist. Diese arterioläre Vasokonstriktion führt unter physiologischen Hb-
Konzentrationen zu einem Abfall des HZV, zu einer Abnahme des koronaren
Blutflusses und einer Einschränkung der funktionellen Kapillardichte. Auf diese
Weise wird das Gewebe-Sauerstoffangebot durch hyperoxische Beatmung in
bestimmten Situationen durch eine weitere Einschränkung der nutritiven
Organversorgung sogar verschlechtert. Hingegen wird während
normovolämischer Hämodilution die hyperoxische Vasokonstriktion durch die
dilutionsbedingte Vasodilatation antagonisiert und die Gewebeoxygenierung
verbessert.
Aus den präsentierten Daten kann geschlossen werden, dass die Beatmung mit
reinem O2 bei hypoxischer Hypoxämie (d.h. bei normalen Hkt-Werten) aufgrund
der zusätzlichen Kompromittierung der Mikrozirkulation infolge hyperoxischer
Vasokonstriktion nicht zwangsläufig zu der intendierten Verbesserung von
Gewebeoxygenierung und Organfunktion führt.
Die klinische Relevanz dieser Ergebnisse muss in künftigen Studien noch
weiter geklärt werden.
Numerical simulation of flow, H₂SO₄ cycle and new particle formation in the CERN CLOUD chamber

To study the effect of galactic cosmic rays on aerosols and clouds, the Cosmic Leaving OUtdoor Droplets (CLOUD) project was established. Experiments are carried out at a 26 m3 tank at CERN (Switzerland). In the experiments, the effect of ionising particle radiation on H2SO4 particle formation and growth is investigated. To evaluate the experimental configuration, the experiment was simulated using a coupled multidimensional CFD – particle model (CLOUD-FPM). In the model the coupled fields of gas/vapour species, temperature, flow velocity and particle properties were computed to investigate the tank's mixing state and mixing times. Simulation results show that the mixing state of the tank's contents largely depends on the characteristics of the mixing fans and a 1-fan configuration, as used in first experiments, may not be sufficient to ensure a homogeneously mixed chamber. To mix the tank properly, 2 fans are necessary. The 1/e response times for instantaneous changes of wall temperature and saturation ratio inside the chamber were found to be in the order of few minutes. Particle nucleation and growth was also simulated and particle number size distribution properties of the freshly nucleated particles (particle number, mean size, standard deviation of the assumed log-normal distribution) were found to be mixed over the tank's volume similar to the gas species.
Calibration of TCCON column-averaged CO₂: the first aircraft campaign over European TCCON sites

The Total Carbon Column Observing Network (TCCON) is a ground-based network of Fourier Transform Spectrometer (FTS) sites around the globe, where the column abundances of CO2, CH4, N2O, CO and O2 are measured. CO2 is constrained with a precision better than 0.25% (1-σ). To achieve a similarly high accuracy, calibration to World Meteorological Organization (WMO) standards is required. This paper introduces the first aircraft calibration campaign of five European TCCON sites and a mobile FTS instrument. A series of WMO standards in-situ profiles were obtained over European TCCON sites via aircraft and compared with retrievals of CO2 column amounts from the TCCON instruments. The results of the campaign show that the FTS measurements are consistently biased 1.1% ± 0.2% low with respect to WMO standards, in agreement with previous TCCON calibration campaigns. The standard a priori profile for the TCCON FTS retrievals is shown to not add a bias. The same calibration factor is generated using aircraft profiles as a priori and with the TCCON standard a priori. With a calibration to WMO standards, the highly precise TCCON CO2 measurements of total column concentrations provide a suitable database for the calibration and validation of nadir-viewing satellites

Globally, tropical forest soils represent the second largest source of N2O and NO. However, there is still considerable uncertainty on the spatial variability and soil properties controlling N trace gas emission. To investigate how soil properties affect N2O and NO emission, we carried out an incubation experiment with soils from 31 locations in the Nyungwe tropical mountain forest in southwestern Rwanda. All soils were incubated at three different moisture levels (50, 70 and 90% water filled pore space (WFPS)) at 17 °C. Nitrous oxide emission varied between 4.5 and 400 μg N m−2 h−1, while NO emission varied from 6.6 to 265 μg N m−2 h−1. Mean N2O emission at different moisture levels was 46.5 ± 11.1 (50% WFPS), 71.7 ± 11.5 (70% WFPS) and 98.8 ± 16.4 (90% WFPS) μg N m−2 h−1, while mean NO emission was 69.3 ± 9.3 (50% WFPS), 47.1 ± 5.8 (70% WFPS) and 36.1 ± 4.2 (90% WFPS) μg N m−2 h−1. The latter suggests that climate (i.e. dry vs. wet season) controls N2O and NO emissions. Positive correlations with soil carbon and nitrogen indicate a biological control over N2O and NO production. But interestingly N2O and NO emissions also showed a negative correlation (only N2O) with soil pH and a positive correlation with free iron. The latter suggest that chemo-denitrification might, at least for N2O, be an important production pathway. In conclusion improved understanding and process based modeling of N trace gas emission from tropical forests will not only benefit from better spatial explicit trace gas emission and basic soil property monitoring, but also by differentiating between biological and chemical pathways for N trace gas formation.
Zum virologischen Nachweis einer akuten Influenza und zur Überprüfung des Immunstatus steht eine Vielzahl von Untersuchungsmethoden zur Verfügung. Bei Verdacht auf eine Influenzavirusinfektion liefert der Rachenabstrich das geeignete Untersuchungsmaterial. Das tiefe Nasopharynxaspirat ist etwas sensitiver, Sputum etwas weniger ergiebig. Die RT-PCR ermöglicht in 1–2 h nach Materialeingang ein sensitives und spezifisches Ergebnis. Typen, Subtypen und Driftvarianten lassen sich durch geeignete Primersonden, die kommerziell zur Verfügung stehen, einwandfrei identifizieren. Demgegenüber ist die Zellkultur-gestützte Virusisolierung zeitaufwendiger und stärker abhängig von einer sachgerechten Materialgewinnung und –überbringung (Kühlkette). PCR und Virusanzüchtung ermöglichen die geno- bzw. phänotypische Testung auf Therapieresistenzen. Der Antigentest ist eine einfache (bed-side) Schnellmethode. Seine Spezifität ist gut, die Sensitivität limitiert; daher kann der Antigentest nicht zur individuellen Ausschlussdiagnose eingesetzt werden. Influenzavirusspezifische Antikörper erscheinen im Blut erst in der zweiten Krankheitswoche. Die Serodiagnostik erfolgt typenspezifisch mit Komplementbindungsreaktion (KBR), IFT und ELISA über eine signifikante Titerbewegung oder den Nachweis von IgA-Antikörpern. IgG-spezifische IFT und ELISA Methoden geben Auskunft über die Influenzavirus-typspezifische Durchseuchung. Die klinisch relevantere subtypen- und variantenspezifische Influenzavirusimmunität wird mit dem HHT oder NT gemessen.
Die Labordiagnose einer Infektionskrankheit beruht auf dem Nachweis des Infektionserregers oder der spezifischen Immunreaktion unter Berücksichtigung der klinischen Plausibilität. Biologische Testverfahren wie der Zellkulturversuch erbringen nur näherungsweise ein quantitatives Ergebnis und sind mit einer relativ großen Streuung behaftet. Das gilt auch für Antikörperassays, soweit sie über ein biologisches Testsignal abgelesen werden (CPE, Agglutination, Komplementverbrauch). Moderne serologische und molekularbiologische Untersuchungsmethoden der Virologie werden i. d. R. über ein physikochemisches Testssignal abgelesen und quantitativ ausgewertet. Dadurch gelingt die nationale und internationale Standardisierung, die sich in Ringversuchen gut überprüfen lässt. Aus biologischen Gründen ist meist eine log. Ergebnisberechnung angezeigt, was für „ signifikante “ Unterschiede in Verlaufsuntersuchungen zu berücksichtigen ist: Da sowohl Infektion als auch Immunreaktion dynamische Prozesse darstellen, können Normalwerte in der virologischen Labordiagnostik nur restriktiv definiert werden. Ihre Ergebnisse sind mehr oder minder individuell interpretationsbedürftig.
The phenomenon of magnetism is a pure quantum effect and has been studied since the beginning of civilization. The practical use of magnetic materials for technical purposes was well established in the 19th century; still nowadays there is no lack of new high-tech applications based on magnetism for example in information technology to store and process data. This thesis does not focus on the development of new applications of magnetism in technology, nor enhancement of known fields of application. Instead, the intention is to use a quantum theory of magnetism for obtaining new insights on physical effects that accompany the phenomenon of magnetism. Therefore three different model systems, each of which are believed to describe a class of real compounds, are considered. Starting from the idea that magnetism can be understood by use of the so-called Heisenberg model that microscopically characterizes the interaction between localized magnetic moments, we restrict ourselves to the case where a long-range magnetic order is present. In order to deduce consequences resulting from this microscopic picture we use the spin-wave theory that is introduced in the first chapter. Central objects of this theory are the magnons which are elementary quantum excitations in ordered magnets. An application of these mathematical techniques to a model that describes an antiferromagnet in an external magnetic field is presented in the second chapter. Quantities like the spin-wave velocity and the damping of magnons are calculated using a Hermitian operator approach in the framework of spin-wave theory. A strong renormalization of the magnetic excitations arises because the symmetry of the system is reduced due to the external magnetic field. In the second model system, that describes thin films of a ferromagnet, concepts of classical physics meet quantum physics: The magnetic dipole-dipole interaction that is also known in everyday life from the magnetic forces between magnets and was initially formulated in the theory of electromagnetism, is included in the microscopic model. Having a special compound in mind where the magnetic excitations are directly accessible in experiments, the energy dispersions of magnon modes in thin-film ferromagnets are deduced. Our approach is essentially a basis for further investigations beyond this thesis to describe strong correlations and condensation of magnons. A recent realization of data processing devices with spin waves puts the understanding of physical processes in these ferromagnetic films in the focus of upcoming research. The third model system brings in the so-called frustration where the interactions between the spins are such that the total energy cannot be minimized by an appropriate alignment of the magnetic moments in the classical picture. In the simplest case this appears because the antiferromagnetically coupled spins are located on a triangular lattice. This situation will lead to strong quantum fluctuations which make this model system interesting. Finally the overall symmetry is reduced by inclusion of spin anisotropies and an external magnetic field. Instead of focusing on the properties of the magnetic excitations, the effect of the magnetic field on the properties of the lattice vibrations is subject to the investigation. This is interesting because the characteristics of lattice vibrations can be measured experimentally using the supersonic technique.

Koreferenzielle Pro-Formen im Deutschen und Italienischen : Analyse von Korpora gesprochener Sprache

Gegenstand dieses Beitrags ist die vergleichende Untersuchung von deutschen und italienischen verweisenden Pronomen. Für das Deutsche wird die Alternanz zwischen der/die/ das (im Folgenden d-Pronomen; vgl. Ahrenholz 2007 und Wiltschko 1998) und Personalpronomen betrachtet. Für das Italienische werden Demonstrativ- und Personalpronomen (freie und klitische) untersucht. In vorherigen Forschungsarbeiten wurde auf semantische, syntaktische und pragmatische Bedingungen hingewiesen, die das Vorkommen von d-Pronomen anstatt anderer Pronominalformen wie z.B. den Personalpronomen im Deutschen stark begünstigen (vgl. dazu Ravetto 2009). Ziel dieser Arbeit ist, zu überprüfen, ob in den zwei Vergleichssprachen ähnliche Tendenzen bzw. Bedingungen zu beobachten sind, die mit der Wahl der jeweiligen verweisenden Form verknüpft sind.
In der vorliegenden Arbeit konzentrierte ich mich auf mediterrane wirbellose Tierarten, welche sich als Konsequenz ihrer Lebensweise nur schlecht ausbreiten können. Nichtsdestotrotz haben es Süßwasserkrabben der Gattung Potamon und Landschnecken der Gattung Tudorella geschafft, große Gebiete zu besiedeln, die heute durch das Mittelmeer getrennt sind. Für beide Gruppen wurde spekuliert, dass Menschen an ihrer Ausbreitung beteiligt waren. Es war mein Ziel die biogeographischen Muster dieser beiden Gattungen zu analysieren und abzuschätzen, ob Menschen tatsächlich Vektoren ihrer Ausbreitung waren. Meine Analysen fanden auf drei Ebenen statt: Taxonomie, Gattung und Art.
Background: Taxonomy or biological systematics is the basic scientific discipline of biology, postulating hypotheses of identity and relationships, on which all other natural sciences dealing with organisms relies. However, the scientific contributions of taxonomists have been largely neglected when using species names in scientific publications by not citing the authority on which they are based.
Discussion: Consequences of this neglect is reduced recognition of the importance of taxonomy, which in turn results in diminished funding, lower interest from journals in publishing taxonomic research, and a reduced number of young scientists entering the field. This has lead to the so-called taxonomic impediment at a time when biodiversity studies are of critical importance.
Here we emphasize a practical and obvious solution to this dilemma. We propose that whenever a species name is used, the author(s) of the species hypothesis be included and the original literature source cited, including taxonomic revisions and identification literature - nothing more than what is done for every other hypothesis or assumption included in a scientific publication. In addition, we postulate that journals primarily publishing taxonomic studies should be indexed in ISISM.
Summary: The proposal outlined above would make visible the true contribution of taxonomists within the scientific community, and would provide a more accurate assessment for funding agencies impact and importance of taxonomy, and help in the recruitment of young scientists into the field, thus helping to alleviate the taxonomic impediment. In addition, it would also make much of the biological literature more robust by reducing or alleviating taxonomic uncertainty.

Forest fragmentation and selective logging are two main drivers of global environmental change and modify biodiversity and environmental conditions in many tropical forests. The consequences of these changes for the functioning of tropical forest ecosystems have rarely been explored in a comprehensive approach. In a Kenyan rainforest, we studied six animal-mediated ecosystem processes and recorded species richness and community composition of all animal taxa involved in these processes. We used linear models and a formal meta-analysis to test whether forest fragmentation and selective logging affected ecosystem processes and biodiversity and used structural equation models to disentangle direct from biodiversity-related indirect effects of human disturbance on multiple ecosystem processes. Fragmentation increased decomposition and reduced antbird predation, while selective logging consistently increased pollination, seed dispersal and army-ant raiding. Fragmentation modified species richness or community composition of five taxa, whereas selective logging did not affect any component of biodiversity. Changes in the abundance of functionally important species were related to lower predation by antbirds and higher decomposition rates in small forest fragments. The positive effects of selective logging on bee pollination, bird seed dispersal and army-ant raiding were direct, i.e. not related to changes in biodiversity, and were probably due to behavioural changes of these highly mobile animal taxa. We conclude that animal-mediated ecosystem processes respond in distinct ways to different types of human disturbance in Kakamega Forest. Our findings suggest that forest fragmentation affects ecosystem processes indirectly by changes in biodiversity, whereas selective logging influences processes directly by modifying local environmental conditions and resource distributions. The positive to neutral effects of selective logging on ecosystem processes show that the functionality of tropical forests can be maintained in moderately disturbed forest fragments. Conservation concepts for tropical forests should thus include not only remaining pristine forests but also functionally viable forest remnants.
Background: Fragmented distribution ranges of species with little active dispersal capacity raise the question about their place of origin and the processes and timing of either range fragmentation or dispersal. The peculiar distribution of the land snail Tudorella sulcata s. str. in Southern France, Sardinia and Algeria is such a challenging case. Methodology: Statistical phylogeographic analyses with mitochondrial COI and nuclear hsp70 haplotypes were used to answer the questions of the species' origin, sequence and timing of dispersal. The origin of the species was on Sardinia. Starting from there, a first expansion to Algeria and then to France took place. Abiotic and zoochorous dispersal could be excluded by considering the species' life style, leaving only anthropogenic translocation as parsimonious explanation. The geographic expansion could be dated to approximately 8,000 years before present with a 95% confidence interval of 10,000 to 3,000 years before present. Conclusions: This period coincides with the Neolithic expansion in the Western Mediterranean, suggesting a role of these settlers as vectors. Our findings thus propose that non-domesticated animals and plants may give hints on the direction and timing of early human expansion routes.
´In die vorliegende klinische Studie wurden 30 Patienten mit einer
nachgewiesenen Erkrankung an rheumatoider Arthritis (RA) und 30 Probanden
ohne Erkrankung an RA einbezogen. Eine Differenzierung innerhalb der
Patientengruppe erfolgte in Bezug auf die Dauer der Erkrankung. Je 15
Patienten, die kürzer bzw. länger als acht Jahre an RA erkrankt waren, wurden
untersucht. Eine Selektion bzgl. des Bestehens einer kraniomandibulären
Dysfunktion (CMD) wurde nicht getroffen. Ziel der Studie war es, die Probanden
und RA-Patienten auf das Vorhandensein von CMD und insbesondere bzgl.
morphologischer Veränderungen der Kiefergelenke zu untersuchen und
vergleichend zu bewerten.
Die Untersuchung der Studienteilnehmer erfolgte anhand des klinischen
Funktionsstatus der Deutschen Gesellschaft für Zahn-, Mund- und
Kieferheilkunde sowie der manuellen Funktionsanalyse nach BUMANN und
GROOT LANDEWEER. Die Ergebnisse der klinischen Funktionsanalyse
wurden mittels des anamnestischen und klinischen Dysfunktionsindexes nach
HELKIMO zusammengefasst und graduiert. Zudem fand eine
magnetresonanztomographische Bewertung (MRT) der Kiefergelenke (n=120)
ohne Kontrastmittelanwendung statt. Zur Erstellung der Aufnahmen kamen
Spinecho-Sequenzen unter Verwendung der T1-, Protonendichte- bzw. T2-
Wichtung mit Fettunterdrückung zum Einsatz. Die Beurteilung erfolgte anhand
eines standardisierten Untersuchungsbogens zur Auswertung von MRTAufnahmen.
Die Kiefergelenke wurden jeweils seitenbezogen bewertet
(rechtsseitig (R)/linksseitig (L)). Die Ergebnisse der Funktionsanalyse ergaben
statistisch signifikante Unterschiede im Vergleich beider Gruppen in Bezug auf
das Vorhandensein von Reibegeräuschen des Kiefergelenks (Probanden 0%,
Patienten 30% (R)/ 33% (L)), bzgl. uni- oder bilateraler Empfindlichkeiten bei
Palpation der Kaumuskulatur (Probanden 37%, Patienten 93%) sowie des
geringeren Ausmaßes der aktiven Mundöffnung bei RA-Patienten. Sowohl der
klinische als auch der anamnestische Dysfunktionsindex nach HELKIMO
belegten die signifikanten Unterschiede im Vergleich beider Gruppen zugunsten
größerer Werte bei den RA-Patienten. Die Auswertung der MRT-Aufnahmen
wies hinsichtlich der Makromorphologie des Kondylus einen signifikanten Unterschied in Bezug auf den Parameter „Deformation“ (Probanden 0%,
Patienten 40% (R)/43% (L)) auf. Mikromorphologische Veränderungen der
Kompakta des Kondylus (Osteophytenbildungen (Probanden 16% (R)/10% (L),
Patienten 67% (R)/80% (L)) und Erosionen (Probanden 6% (R)/0% (L),
Patienten 47% (R)/40% (L)) sowie der kondylären Spongiosa (Probanden 0%,
Patienten 53% (R)/50% (L)) existierten signifikant häufiger innerhalb der RAGruppe.
Die Kondylus-Diskus-Relation (mundgeschlossen/parasagittal)
differierte ausgehend von einer regelrechten Position (Probanden 67% (R)/83%
(L), Patienten 33% (R)/27% (L)) häufiger zu einer partiellen anterioren
Diskusverlagerung (DV) mit Reposition (Probanden 30% (R)/10% (L), Patienten
33% (R)/37% (L)) sowie zu einer totalen anterioren DV ohne Reposition
(Probanden 0% (R)/3% (L), Patienten 17% (R)/20% (L)) innerhalb der RAGruppe.
Signalreiche Anteile in der bilaminären Zone bzw. im Gelenkspalt, die
auf intraartikuläre Flüssigkeitsansammlungen hinweisen, traten signifikant
häufiger innerhalb der RA-Gruppe auf (Probanden 3% (R)/7% (L), Patienten
80% (R)/90% (L)). Ein Zusammenhang zwischen dem Vorhandensein von
Reibegeräuschen und spezifischen osteoarthrotischen Veränderungen im MRTBefund
sowie ein Zusammenhang zwischen einer klinisch eingeschränkten
Mundöffnung und einer im MRT-Befund nachweisbaren totalen anterioren DV
ohne Reposition konnten jeweils statistisch nachgewiesen werden. Ein
Zusammenhang zwischen einer klinischen Palpationsempfindlichkeit
(intraaurikulär), einer Schmerzhaftigkeit bei passiver Kompression bzw. bei
ventrokaudaler Translation mit dem Vorliegen von intraartikulären
Flüssigkeitsansammlungen im MRT-Befund ließ sich jeweils statistisch nicht
bestätigen. Des Weiteren wurde aufgezeigt, dass die Dauer der Erkrankung an
RA nicht mit den Dysfunktionsindizes nach HELKIMO und ebenso nicht mit
ausgewählten MRT-Befunden (z.B. Deformation des Kondylus,
Osteophytenbildungen/Erosionen der Kompakta des Kondylus, degenerative
Veränderungen der Spongiosa des Kondylus, Deformation des Diskus und
Vorliegen einer totalen anterioren DV ohne Reposition) korreliert.
Die Ergebnisse dieser Studie weisen darauf hin, dass ein an RA erkrankter
Patient mit einer beträchtlichen Wahrscheinlichkeit Dysfunktionen sowie
morphologische Veränderungen im Bereich der Kiefergelenke entwickeln kann,
die eine frühzeitige Untersuchung dieses Gelenkes als sinnvoll erachten lassen.
