We study the charmonium coherent photoproduction and hadroproduction consistently with modifications from both cold and hot nuclear matters. The strong electromagnetic fields from fast moving nucleus interact with the other target nucleus, producing abundant charmonium in the extremely low transverse momentum region pT<0.1 GeV/c. This results in significative enhancement of J/ψ nuclear modification factor in semi-central and peripheral collisions. In the middle pT region such as pT<3∼5 GeV/c, J/ψ final yield is dominated by the combination process of single charm and anti-charm quarks moving in the deconfined matter, c+c¯→J/ψ+g. In the higher pT region, J/ψ production are mainly from parton initial hard scatterings at the beginning of nucleus–nucleus collisions and decay of B hadrons. We include all of these production mechanisms and explain the experimental data well in different colliding centralities and transverse momentum regions.
Mobilization of hematopoietic stem cells (HSCs) from the bone marrow to the peripheral blood is a complex mechanism that involves adhesive and chemotactic interactions of HSCs as well as their bone marrow microenvironment. In addition to a number of non-genetic factors, genetic susceptibilities also contribute to the mobilization outcome. Identification of genetic factors associated with HSC yield is important to better understand the mechanism behind HSC mobilization. In the present study, we enrolled 148 Korean participants (56 healthy donors and 92 patients) undergoing HSC mobilization for allogeneic or autologous HSC transplantation. Among a total of 53 polymorphisms in 33 candidate genes, one polymorphism (rs11264422) in relaxin/insulin-like family peptide receptor 4 (RXFP4) gene was significantly associated with a higher HSC yield after mobilization in Koreans. However, in a set of 101 Europeans, no association was found between circulating CD34+ cell counts and rs11264422 genotype. Therefore, we suggest that the ethnic differences in subjects’ genetic background may be related to HSC mobilization. In conclusion, the relaxin—relaxin receptor axis may play an important role in HSC mobilization. We believe that the results of the current study could provide new insights for therapies that use relaxin and HSC populations, as well as a better understanding of HSC regulation and mobilization at the molecular level.
No presente artigo analisamos a ideia de solidariedade na teoria da justiça de Axel Honneth. Honneth em sua obra atualiza a teoria hegeliana da eticidade e concebe a existência de esferas de liberdade, sendo que uma delas é a liberdade social, na qual se situam relações pessoais, mercado e instituições políticas. Em O Direito da Liberdade Honneth desenvolve uma teoria em que busca analisar os problemas de desenvolvimento da democracia nas sociedades contemporâneas, e erige a solidariedade como um dos pressupostos normativos para a construção de uma democracia virtuosa, ancorada na participação cidadã.
O presente estudo tem como objetivo investigar a ideia de reconhecimento jurídico na teoria de Axel Honneth, o que se dará mediante análise da obra Luta por Reconhecimento. Honneth, ancorado nas teorias de Hegel e Mead, estabelece o papel do direito como esfera de reconhecimento individual e seu potencial de asseguramento do autorrespeito. Empreende-se uma reconstrução da teoria honnethiana no atinente aos papéis desempenhados pelo direito na teoria da Luta por Reconhecimento de Axel Honneth, para em seguida, a partir da leitura que o autor desenvolve da teoria de Thomas Marshall, analisar o papel desempenhado pelos direitos subjetivos fundamentais como medium de sedimentação e ampliação de novas formas de reconhecimento e cidadania.
Information theory provides a formal framework within which information processing and its disorders can be described. However, information theory has rarely been applied to modeling aspects of the cognitive neuroscience of schizophrenia. The goal of this article is to highlight the benefits of an approach based on information theory, including its recent extensions, for understanding several disrupted neural goal functions as well as related cognitive and symptomatic phenomena in schizophrenia. We begin by demonstrating that foundational concepts from information theory—such as Shannon information, entropy, data compression, block coding, and strategies to increase the signal-to-noise ratio—can be used to provide novel understandings of cognitive impairments in schizophrenia and metrics to evaluate their integrity. We then describe more recent developments in information theory, including the concepts of infomax, coherent infomax, and coding with synergy, to demonstrate how these can be used to develop computational models of schizophrenia-related failures in the tuning of sensory neurons, gain control, perceptual organization, thought organization, selective attention, context processing, predictive coding, and cognitive control. Throughout, we demonstrate how disordered mechanisms may explain both perceptual/cognitive changes and symptom emergence in schizophrenia. Finally, we demonstrate that there is consistency between some information-theoretic concepts and recent discoveries in neurobiology, especially involving the existence of distinct sites for the accumulation of driving input and contextual information prior to their interaction. This convergence can be used to guide future theory, experiment, and treatment development.
Bromus racemosus L. is a rather rare grass species of moist meadows. It has strongly decreased in the course of the 20th century due to intensification of agricultural grassland management, and is therefore included in Red Lists of several European countries. Its winter annual life-cycle is remarkable for a species of permanent grasslands.
The aim of this study is to determine the habitat preference and optimal management of B. racemosus in the Netherlands and surrounding countries. Vegetation, soil and hydrological data from 28 sites in the Netherlands have been compared with B. racemosus cover, and with vegetation data from surrounding countries. The results indicate that B. racemosus is characteristic of Molinio-Arrhenatheretea meadows with good mineralisation and aftermath grazing. The optimum lies in grasslands of the alliance Alopecurion pratensis (Deschampsion cespitosae), but the species ranges from wetter Calthion palustris meadows to drier Arrhenatherion elatioris and Cynosurion cristati grasslands. It prefers intermediate nutrient levels and hydrological conditions (mesic sites), but within this range the highest cover is found in relatively nutrient rich and dry sites. Because of the absence of a seedbank and a low dispersal capability, B. racemosus is vulnerable to changes in grassland management. A management of mowing after 15 June and aftermath grazing is most suitable, since it enables fruit ripening and the maintenance of an open sward, needed for germination and development. The risk of extinction is likely to be higher in flat polders than in floodplain sites with natural relief, where the species may shift between belts in different years.
Im Jahr 1926 erschien in der 'Revista de Occidente', einer von José Ortega y Gasset in Madrid herausgegebenen Zeitschrift mit kulturpolitischer und philosophischer Ausrichtung, eine merkwürdige Publikation unter dem rätselhaften Titel "Cancionero apócrifo de Abel Martín. Recopilación y estudio de Juan de Mairena." Auf den ersten Blick scheint es so, als fungiere Antonio Machado hier lediglich als Herausgeber einer von Juan de Mairena kompilierten Sammlung, welche wiederum der Autorschaft eines Abel Martín zugeschrieben wird. Trotz der rätselhaften doppelten Herausgeberfiktion hatte das damalige Lesepublikum offenbar keine Probleme damit, Antonio Machado als den eigentlichen Autor der genannten "apokryphen" Prosa- und Gedichttexte zu identifizieren. Denn die Namen Juan de Mairena und Abel Martín bezeichnen nicht etwa reale Dichterpersönlichkeiten der damaligen Epoche oder der spanischen Literatur- geschichte, sondern sie sind – wie die damaligen Leser durch Ausschlussverfahren selbst erraten konnten – Konstrukte von Machados Phantasie, fiktive Autorprojektionen, deren Erfindung und Gestaltung er mit erstaunlichem gedanklichen Aufwand und Akribie betrieben hatte. Abel Martín und sein 'Schüler' Mairena gehören zu einem ganzen Spektrum von imaginierten Dichtern und Philosophen, deren fingierte Werke ihr Erfinder Machado als "apokryph" bezeichnete. Die bemerkenswerte Praktik, eigene Hervorbringungen, Lyrik und Prosa, nicht unter dem eigenen Namen zu publizieren, sondern einem Kreis fingierter Autorpersönlichkeiten zuzuordnen, hebt sich von dem gewöhnlichen Gebrauch von Pseudonymen, von künstlerischen Decknamen, deutlich ab. Zumal der Autor in jenem Zeitraum in den 1910er und 1920er Jahren keineswegs mehr ein Unbekannter war.
Dem Apokryphen einen Themenschwerpunkt im Rahmen einer komparatistischen Zeitschrift zu widmen ist eine Entscheidung, die auf den ersten Blick erstaunen mag. Ist doch das Wort "apokryph" ein in der heutigen Alltagssprache wenig geläufiger Begriff, der, auf das Register der Theologie verweisend, speziellen Fragen und fachwissenschaftlichen Debatten der Religionsgeschichte vorbehalten zu sein scheint. Wenn wir gleichwohl Begriff und Phänomen des Apokryphen als Schwerpunktthema des Jahrbuchs 'Komparatistik' zur Diskussion stellen wollen, so deshalb, weil der damit angesprochene Sachverhalt von Prozessen der Auswahl, Marginalisierung und Ausgrenzung im Rahmen kultureller und textueller Überlieferung ein Problemfeld markiert, das im Bereich der Literaturgeschichte vielfältige Resonanzen und Parallelen findet und in systematischer Hinsicht über Verfahren und Mechanismen literarischer Traditionsbildung Aufschluss zu geben verspricht. Weit davon entfernt, nur ein spezialdiskursives Sonderphänomen zu sein, erweist sich das Apokryphe bei näherem Hinsehen als eine Figur, an der sich grundsätzliche Fragen der Formation von Text- und Wissensbeständen, der literarischen Autorität und Kanonbildung beobachten lassen. Apokryphe Texte, so ließe sich in einer vorläufigen, im Folgenden noch zu präzisierenden Beschreibung formulieren, sind Texte, die sich am Rande der großen Traditionen religiöser und kultureller Bewegungen situieren. Es sind Texte oder Textensembles, die es, wie sich im Rückblick bemerken lässt, nicht geschafft haben, in das Inventar jener Schriften aufgenommen zu werden, die als anerkannt, bewahrenswert oder wahr gelten. Als Einstieg bietet es sich an, zunächst von den Bedeutungsdimensionen des Ausdrucks apokryph auszugehen, die dieser im Kontext der religionsgeschichtlichen Tradition angenommen hat und der für das heute übliche Verständnis des Begriffs bestimmend ist. Der religiöse bzw. theologische Begriffsgebrauch greift auf das altgriechische Wort ἀπόκρυφος (apókryphos) zurück, um dieses vor allem in einer seiner beiden Bedeutungsschichten zum Einsatz zu bringen, nämlich als Bezeichnung eines Gegenstands bzw. Textes, der als 'unecht', 'zweifelhaft', 'nicht-authentisch' zu gelten habe. Der Begriff apokryph führt hier also eine Unterscheidung ein zwischen dem, was als gesichert, gültig und autorisiert anzusehen ist und dem, was solche Geltung zu besitzen nicht oder nur unter dem Vorbehalt des Zweifelhaften beanspruchen kann. Bei dem Begriffspaar gültig vs. apokryph haben wir es also mit einer Figur der Verknappung zu tun, die ein gegebenes Textensemble oder eine kulturelle Überlieferung einem mitunter radikalen Verfahren der Auswahl und Reduktion unterzieht.
The yeast bc1 complex (complex III) and cytochrome oxidase (complex IV) are mosaics of core subunits encoded by the mitochondrial genome and additional nuclear-encoded proteins imported from the cytosol. Both complexes build in the mitochondrial inner membrane various supramolecular assemblies. The formation of the individual complexes and their supercomplexes depends on the activity of dedicated assembly factors. We identified a so far uncharacterized mitochondrial protein (open reading frame YDR381C-A) as an important assembly factor for complex III, complex IV, and their supercomplexes. Therefore, we named this protein Cox interacting (Coi) 1. Deletion of COI1 results in decreased respiratory growth, reduced membrane potential, and hampered respiration, as well as slow fermentative growth at low temperature. In addition, coi1Δ cells harbour reduced steady-state levels of subunits of complexes III and IV as well as of the assembled complexes and supercomplexes. Interaction of Coi1 with respiratory chain subunits seems transient, as it appears to be a stoichiometric subunit neither of complex III nor of complex IV. Collectively, this work identifies a novel protein that plays a role in the assembly of the mitochondrial respiratory chain.
Most event studies rely on cumulative abnormal returns, measured as percentage changes in stock prices, as their dependent variable. Stock price reflects the value of the operating business plus non-operating assets minus debt. Yet, many events, in particular in marketing, only influence the value of the operating business, but not non-operating assets and debt. For these cases, the authors argue that the cumulative abnormal return on the operating business, defined as the ratio between the cumulative abnormal return on stock price and the firm-specific leverage effect, is a more appropriate dependent variable. Ignoring the differences in firm-specific leverage effects inflates the impact of observations pertaining to firms with large debt and deflates those pertaining to firms with large non-operating assets. Observations of firms with high debt receive several times the weight attributed to firms with low debt. A simulation study and the reanalysis of three previously published marketing event studies shows that ignoring the firm-specific leverage effects influences an event study's results in unpredictable ways.
The physical housing environment is important to facilitate activities of daily living (ADL) for older people. A hindering environment may lead to ADL dependence and thus increase the need for home services, which is individually restricting and a growing societal burden. This study presents simulations of policy changes with regard to housing accessibility that estimates the potential impact specifically on instrumental activities of daily living (I-ADL), usage of home services, and related costs. The models integrate empirical data to test the hypothesis that a policy providing funding to remove the five most severe environmental barriers in the homes of older people who are at risk of developing dependence in I-ADL, can maintain independence and reduce the need for home services. In addition to official statistics from state agencies in Sweden and Germany, we utilized published results from the ENABLE-AGE and other scientific studies to generate the simulations. The simulations predicted that new policies that remove potentially hindering housing features would improve I-ADL performance among older people and reduce the need for home services. Our findings suggest that a policy change can contribute to positive effects with regard to I-ADL independence among older people and to a reduction of societal burden.
Vegetation patterns of floodplain forests are highly variable across different habitats in European regions. Their plant communities have been well described from the phytosociological point of view, but plant species richness and composition patterns and their underlying environmental factors are still insufficiently known. Sixty-one vegetation plots of black alder-dominated floodplain forests were sampled in central Slovakia in order to find main environmental predictors affecting floristic diversity of their vegetation. For each vegetation plot with a constant size (400m2), vascular plant species and a set of topographic, climatic, soil physical and chemical characteristics were recorded. A generalized linear model was applied to explain relevance of environmental factors on changes of species richness, whereas the relationship between species composition and explanatory variables was tested using ordination methods. Main gradients of species compositional variation were soil moisture, light, elevation and soil chemistry-related variables. Vascular species richness of plots varied between 19 and 59 (mean 38). Herb-layer species richness was positively related to the soil pH, stream power index and negatively to the concentration of soil iron. These linear trends were accompanied by a hump-shaped response to sand content and a U-shaped response to elevation.
Sex differences in the relationship between conduct disorder and cortical structure in adolescents

Objective: Previous studies have reported reduced cortical thickness and surface area and altered gyrification in frontal and temporal regions in adolescents with conduct disorder (CD). Although there is evidence that the clinical phenotype of CD differs between males and females, no studies have examined whether such sex differences extend to cortical and subcortical structure.
Method: As part of a European multisite study (FemNAT-CD), structural magnetic resonance imaging (MRI) data were collected from 48 female and 48 male participants with CD and from 104 sex-, age-, and pubertal-status−matched controls (14–18 years of age). Data were analyzed using surface-based morphometry, testing for effects of sex, diagnosis, and sex-by-diagnosis interactions, while controlling for age, IQ, scan site, and total gray matter volume.
Results: CD was associated with cortical thinning and higher gyrification in ventromedial prefrontal cortex in both sexes. Males with CD showed lower, and females with CD showed higher, supramarginal gyrus cortical thickness compared with controls. Relative to controls, males with CD showed higher gyrification and surface area in superior frontal gyrus, whereas the opposite pattern was seen in females. There were no effects of diagnosis or sex-by-diagnosis interactions on subcortical volumes. Results are discussed with regard to attention-deficit/hyperactivity disorder, depression, and substance abuse comorbidity, medication use, handedness, and CD age of onset.
Conclusion: We found both similarities and differences between males and females in CD–cortical structure associations. This initial evidence that the pathophysiological basis of CD may be partly sex-specific highlights the need to consider sex in future neuroimaging studies and suggests that males and females may require different treatments.
The existence of individual variation in males' motivation to mate remains a conundrum as directional selection should favour high mating frequencies. Balancing selection resulting from (context-dependent) female mate choice could contribute to the maintenance of this behavioural polymorphism. In dichotomous choice tests, mosquitofish (Gambusia holbrooki) females preferred virtual males showing intermediate mating frequencies, reflecting females' tendencies to avoid harassment by highly sexually active males. When tested in the presence of a female shoal—which protects females from male harassment—focal females showed significantly stronger preferences for high sexual activity. A trade-off between (indirect) benefits and (direct) costs of mating with sexually active males probably explains context-dependent female mate choice, as costs depend on the social environment in which females choose their mates. No preference was observed when we tested virgin females, suggesting that the behavioural pattern described here is part of the learned behavioural repertoire of G. holbrooki females.
BACKGROUND: Time-limited, early-life exposures to institutional deprivation are associated with disorders in childhood, but it is unknown whether effects persist into adulthood. We used data from the English and Romanian Adoptees study to assess whether deprivation-associated adverse neurodevelopmental and mental health outcomes persist into young adulthood.
METHODS: The English and Romanian Adoptees study is a longitudinal, natural experiment investigation into the long-term outcomes of individuals who spent from soon after birth to up to 43 months in severe deprivation in Romanian institutions before being adopted into the UK. We used developmentally appropriate standard questionnaires, interviews completed by parents and adoptees, and direct measures of IQ to measure symptoms of autism spectrum disorder, inattention and overactivity, disinhibited social engagement, conduct or emotional problems, and cognitive impairment (IQ score <80) during childhood (ages 6, 11, and 15 years) and in young adulthood (22-25 years). For analysis, Romanian adoptees were split into those who spent less than 6 months in an institution and those who spent more than 6 months in an institution. We used a comparison group of UK adoptees who did not experience deprivation. We used mixed-effects regression models for ordered-categorical outcome variables to compare symptom levels and trends between groups.
FINDINGS: Romanian adoptees who experienced less than 6 months in an institution (n=67 at ages 6 years; n=50 at young adulthood) and UK controls (n=52 at age 6 years; n=39 at young adulthood) had similarly low levels of symptoms across most ages and outcomes. By contrast, Romanian adoptees exposed to more than 6 months in an institution (n=98 at ages 6 years; n=72 at young adulthood) had persistently higher rates than UK controls of symptoms of autism spectrum disorder, disinhibited social engagement, and inattention and overactivity through to young adulthood (pooled p<0·0001 for all). Cognitive impairment in the group who spent more than 6 months in an institution remitted from markedly higher rates at ages 6 years (p=0·0001) and 11 years (p=0·0016) compared with UK controls, to normal rates at young adulthood (p=0·76). By contrast, self-rated emotional symptoms showed a late-onset pattern with minimal differences versus UK controls at ages 11 years (p=0·0449) and 15 years (p=0·17), and then marked increases by young adulthood (p=0·0005), with similar effects seen for parent ratings. The high deprivation group also had a higher proportion of people with low educational achievement (p=0·0195), unemployment (p=0·0124), and mental health service use (p=0·0120, p=0·0032, and p=0·0003 for use when aged <11 years, 11-14 years, and 15-23 years, respectively) than the UK control group. A fifth (n=15) of individuals who spent more than 6 months in an institution were problem-free at all assessments.
INTERPRETATION: Notwithstanding the resilience shown by some adoptees and the adult remission of cognitive impairment, extended early deprivation was associated with long-term deleterious effects on wellbeing that seem insusceptible to years of nurturance and support in adoptive families.

Este artigo apresenta uma discussão sobre a obra recente de Axel Honneth. Começando por uma exposição de seu projeto teórico mais amplo, descrito em O direito da liberdade (2011) como uma teoria da justiça sociologicamente ancorada, o artigo demonstra como Honneth se move da ideia de uma luta por reconhecimento em direção à ideia de intersubjetividade institucional. Este movimento, porém, é acompanhado por críticas que vêm nesta passagem o abandono das ambições críticas de seu modelo e um compromisso reformista com a ordem capitalista de mercado. A fim de responder a estas objeções, Honneth propõe uma reatualização da ideia do socialismo (2015) a partir da noção de liberdade social, concluindo com uma distinção entre duas formas de luta por reconhecimento, uma interna e outra externa – e defendendo as vantagens da primeira.
From reciprocal recognition to a society that is properly 'social' : on Axel Honneth's recent work

This paper addresses Axel Honneth's recent endeavors to defend his theory of justice, broadly described in Freedom's right (2011) as an analysis of society. The paper begins by exposing Honneth's model as a theory of institutional intersubjectivity rather than a theory of the struggle for recognition. This model, however, was subject to criticism due to its supposed acceptance of the capitalistic market economy as a social order. In order to defend it from such objections, Honneth (2016) exposes the normative core of socialist ideals as a version of social freedom. Finally, he presents a distinction between two forms of political intervention: an internal and an external struggle for recognition – and asserts the advantages of the former.
Auf Grundlage einer interviewbasierten Studie zu heterosexuellen Paaren, in denen die Frau das Haupteinkommen verdient, beschäftigt sich der Beitrag mit milieuspezifischen Bewältigungsmustern prekärer Beschäftigungsverhältnisse. Vor dem Hintergrund der Erosion des Ernährermodells werden dabei Transformationen von Männlichkeit in den Blick genommen. Es wird die These entwickelt, dass sich mit dem Selbstverständnis als "Künstler" im hochqualifizierten individualisierten Milieu des urbanen Raums ein spezifisches Bewältigungsmuster von Prekarität herausgebildet hat.
Correlation of lumbar lateral recess stenosis in magnetic resonance imaging and clinical symptoms

Aim: To assess the correlation of lateral recess stenosis (LRS) of lumbar segments L4/5 and L5/S1 and the Oswestry Disability Index (ODI).
Methods: Nine hundred and twenty-seven patients with history of low back pain were included in this uncontrolled study. On magnetic resonance images (MRI) the lateral recesses (LR) at lumbar levels L4/5 and L5/S1 were evaluated and each nerve root was classified into a 4-point grading scale (Grade 0-3) as normal, not deviated, deviated or compressed. Patient symptoms and disability were assessed using ODI. The Spearman’s rank correlation coefficient was used for statistical analysis (P < 0.05).
Results: Approximately half of the LR revealed stenosis (grade 1-3; 52% at level L4/5 and 42% at level L5/S1) with 2.2% and 1.9% respectively reveal a nerve root compression. The ODI score ranged from 0%-91.11% with an arithmetic mean of 34.06% ± 16.89%. We observed a very weak statistically significant positive correlation between ODI and LRS at lumbar levels L4/5 and L5/S1, each bilaterally (L4/5 left: rho < 0.105, P < 0.01; L4/5 right: rho < 0.111, P < 0.01; L5/S1 left: rho 0.128, P < 0.01; L5/S1 right: rho < 0.157, P < 0.001).
Conclusion: Although MRI is the standard imaging tool for diagnosing lumbar spinal stenosis, this study showed only a weak correlation of LRS on MRI and clinical findings. This can be attributed to a number of reasons outlined in this study, underlining that imaging findings alone are not sufficient to establish a reliable diagnosis for patients with LRS.
In Europe, anthroponyms have undergone a complex development, as simple one-word personal names have developed into multi-word structures containing one, two or more personal names and a stable surname. The article predominantly characterizes the development of surnames on the basis of their origin. Attention is also paid to different trends in the development of personal names. The article analyses this phenomenon from various perspectives, using an example of one specific living person’s anthroponyms.
Nerve tissue contains a high density of chemical synapses, about 1 per µm3 in the mammalian cerebral cortex. Thus, even for small blocks of nerve tissue, dense connectomic mapping requires the identification of millions to billions of synapses. While the focus of connectomic data analysis has been on neurite reconstruction, synapse detection becomes limiting when datasets grow in size and dense mapping is required. Here, we report SynEM, a method for automated detection of synapses from conventionally en-bloc stained 3D electron microscopy image stacks. The approach is based on a segmentation of the image data and focuses on classifying borders between neuronal processes as synaptic or non-synaptic. SynEM yields 97% precision and recall in binary cortical connectomes with no user interaction. It scales to large volumes of cortical neuropil, plausibly even whole-brain datasets. SynEM removes the burden of manual synapse annotation for large densely mapped connectomes.
Background: Infections with the hepatitis C virus (HCV) are a global public health problem. Long-term consequences are the development of liver cirrhosis and hepatocellular carcinoma. Newly introduced direct acting antivirals, especially interferon-free regimens, have improved rates of sustained viral response above 90% in most patient groups and allow treating patients who were ineligible for treatment in the past. These new regimens have replaced former treatment and are recommended by current guidelines. However, there is an ongoing discussion on high pharmaceutical prices. Our aim was to assess the long-term cost-effectiveness of treating hepatitis C genotype 1 patients with sofosbuvir/ledipasvir (SOF/LDV) treatment in Germany.
Material and Methods: We used a Markov cohort model to simulate disease progression and assess cost-effectiveness. The model calculates lifetime costs and outcomes (quality-adjusted life years, QALYs) of SOF/LDV and other strategies. Patients were stratified by treatment status (treatment-naive and treatment-experienced) and absence/presence of cirrhosis. Different treatment strategies were compared to prior standard of care. Sensitivity analyses were performed to evaluate model robustness.
Results: Base-case analyses results show that in treatment-naive non-cirrhotic patients treatment with SOF/LDV dominates the prior standard of care (is more effective and less costly). In cirrhotic patients an incremental cost-effectiveness ratio (ICER) of 3,383 €/QALY was estimated. In treatment-experienced patients ICERs were 26,426 €/QALY and 1,397 €/QALY for treatment-naive and treatment-experienced patients, respectively. Robustness of results was confirmed in sensitivity analyses.
Conclusions: Our analysis shows that treatment with SOF/LDV is cost-effective compared to prior standard of care in all patient groups considering international costs per QALY thresholds.
The accurate knowledge of the neutron-induced fission cross-sections of actinides and other isotopes involved in the nuclear fuel cycle is essential for the design of advanced nuclear systems, such as Generation-IV nuclear reactors. Such experimental data can also provide the necessary feedback for the adjustment of nuclear model parameters used in the evaluation process, resulting in the further development of nuclear fission models. In the present work, the 240Pu(n,f) cross-section was measured at CERN's n_TOF facility relative to the well-known 235U(n,f) cross section, over a wide range of neutron energies, from meV to almost MeV, using the time-of-flight technique and a set-up based on Micromegas detectors. This measurement was the first experiment to be performed at n_TOF's new experimental area (EAR-2), which offers a significantly higher neutron flux compared to the already existing experimental area (EAR-1). Preliminary results as well as the experimental procedure, including a description of the facility and the data handling and analysis, are presented.
Tumor cells frequently overexpress heat shock protein 70 (Hsp70) and present it on their cell surface, where it can be recognized by pre‐activated NK cells. In our retrospective study the expression of Hsp70 was determined in relation to tumor‐infiltrating CD56+ NK cells in formalin‐fixed paraffin embedded (FFPE) tumor specimens of patients with SCCHN (N = 145) as potential indicators for survival and disease recurrence. All patients received radical surgery and postoperative cisplatin‐based radiochemotherapy (RCT). In general, Hsp70 expression was stronger, but with variable intensities, in tumor compared to normal tissues. Patients with high Hsp70 expressing tumors (scores 3–4) showed significantly decreased overall survival (OS; p = 0.008), local progression‐free survival (LPFS; p = 0.034) and distant metastases‐free survival (DMFS; p = 0.044), compared to those with low Hsp70 expression (scores 0–2), which remained significant after adjustment for relevant prognostic variables. The adverse prognostic value of a high Hsp70 expression for OS was also observed in patient cohorts with p16‐ (p = 0.001), p53‐ (p = 0.0003) and HPV16 DNA‐negative (p = 0.001) tumors. The absence or low numbers of tumor‐infiltrating CD56+ NK cells also correlated with significantly decreased OS (p = 0.0001), LPFS (p = 0.0009) and DMFS (p = 0.0001). A high Hsp70 expression and low numbers of tumor‐infiltrating NK cells have the highest negative predictive value (p = 0.00004). In summary, a strong Hsp70 expression and low numbers of tumor‐infiltrating NK cells correlate with unfavorable outcome following surgery and RCT in patients with SCCHN, and thus serve as negative prognostic markers.

In bacteria, the regulation of gene expression by cis-acting transcriptional riboswitches located in the 5'-untranslated regions of messenger RNA requires the temporal synchronization of RNA synthesis and ligand binding-dependent conformational refolding. Ligand binding to the aptamer domain of the riboswitch induces premature termination of the mRNA synthesis of ligand-associated genes due to the coupled formation of 3'-structural elements acting as terminators. To date, there has been no high resolution structural description of the concerted process of synthesis and ligand-induced restructuring of the regulatory RNA element. Here, we show that for the guanine-sensing xpt-pbuX riboswitch from Bacillus subtilis, the conformation of the full-length transcripts is static: it exclusively populates the functional off-state but cannot switch to the on-state, regardless of the presence or absence of ligand. We show that only the combined matching of transcription rates and ligand binding enables transcription intermediates to undergo ligand-dependent conformational refolding.
Die Verkleidung des Richters und die Verkleidung der Muslima sind ähnlich, weil sie beide aus einer Abschirmung rühren, die auf gleiche Weise dogmatisch besetzt ist. Auch wenn, zumindest in der deutschen Übersetzung des Koran, mit der Bedeckung der Haare die Keuschheit, Sittsamkeit oder Schamhaftigkeit der Frau und mit der Verkleidung des Richters seine Neutralität symbolisiert oder umgesetzt werden soll, gibt es doch eine Entsprechung, und die liegt in der Abschirmung. Wenn man hijab mit Absperrung oder Verhüllung übersetzt, wie das einige vorschlagen, dann tragen Richter auch einen hijab.
Die gleichgeschlechtliche Ehe wurde vom Gesetzgeber endlich anerkannt, auch das Verfassungsgericht wird dagegen nichts mehr unternehmen können, das hoffe ich zumindest. Wer seine Zeichen und Gesetze dennoch exklusiv halten will, sollte darum darüber nachdenken, auf Alternativbegriffe zur Ehe umzusteigen.

Die eifrigste Bestrebung der Kinder besteht darin, so hat es zumindest Sigmund Freud einmal behauptet, "zu erfahren, was die Eltern miteinander tun, woraus dann die Kinder werden." Was treibt die Kinder in die Welt? Haben die Kinder Juristen als Eltern, dann dürften sie bald eher nach den Gründen als nach dem Treiben fragen.
Wenn Rechtstexte auf literarische Texte treffen, dann treffen Wahrheitsformen aufeinander. Es treffen unterschiedliche Weisen, Wahrheit zu produzieren, aufeinander. Zu den zahlreichen Unterschieden gehören, historisch bedingt, unterschiedliche Stile, die sich um die Objektivierung und "Subjektivierung" der Aussagen bilden. Das fängt bei den banalen "wir" und "man" rechtswissenschaftlicher Texte an, geht über allgemein gehaltene, enthistorisierende und systematisierende Definitionen bis zu einem Fussnotenapparat, der in manchen Rechtstexten beinahe jede Aussage als nachweisbare Aussage absichern soll. Die Rechtswissenschaft, zumal die deutsche, pflegt bei ihren Wahrheitsformen objektivierende Stile, die Literaturwissenschaft tut das nicht, nicht in dem Maße, sie lässt das Subjekt stärker in die Aussage einbrechen. Die Lage zwischen diesen beiden Disziplinen ist allerdings kompliziert, weil wiederum das Recht eine wirksame Subjektivierungsinstanz ist, die eben die Instrumente zur Verfügung stellt, um Aussagen an Subjekte zu binden. Sie hat die Unterschrift und den Urheber erfunden. In dieser Lage kann man also nicht einfach das Subjektive gegen das Objektive ausspielen. Von Anfang aber über das Zusammentreffen von Literatur und Recht in objektivierenden Stilen zu schreiben, schafft eine asymmetrische Ausgangslage, die ich gerne zugunsten einer symmetrischen Ausgangslage umgehen möchte – soweit das möglich ist. ...
We study the correlation between the distributions of the net-charge, net-kaon, net-baryon and net-proton number at hadronization and after the final hadronic decoupling by simulating ultra relativistic heavy ion collisions with the hybrid version of the ultrarelativistic quantum molecular dynamics (UrQMD) model. We find that due to the hadronic rescattering these distributions are not strongly correlated. The calculated change of the correlation, during the hadronic expansion stage, does not support the recent paradigm, namely that the measured final moments of the experimentally observed distributions do give directly the values of those distributions at earlier times, when the system had been closer to the QCD crossover.
Chromosomal translocations - leading to the expression of fusion genes - are well-studied genetic abberrations associated with the development of leukemias. Most of them represent altered transcription factors that affect transcription or epigenetics, while others - like BCR-ABL - are enhancing signaling. BCR-ABL has become the prototype for rational drug design, and drugs like Imatinib and subsequently improved drugs have a great impact on cancer treatments. By contrast, MLL-translocations in acute leukemia patients are hard to treat, display a high relapse rate and the overall survival rate is still very poor. Therefore, new treatment modalities are urgently needed. Based on the molecular insights of the most frequent MLL rearrangements, BET-, DOT1L-, SET- and MEN1/LEDGF-inhibitors have been developed and first clinical studies were initiated. Not all results of these studies have are yet available, however, a first paper reports a failure in the DOT1L-inhibitor study although it was the most promising drug based on literature data. One possible explanation is that all of the above mentioned drugs also target the cognate wildtype proteins. Here, we want to strengthen the fact that efforts should be made to develop drugs or strategies to selectively inhibit only the fusion proteins. Some examples will be given that follow exactly this guideline, and proof-of-concept experiments have already demonstrated their feasibility and effectiveness. Some of the mentioned approaches were using drugs that are already on the market, indicating that there are existing opportunities for the future which should be implemented in future therapy strategies.

Dann stellt sich zugleich die Frage: Was kann man tun, wenn man den Auftritt eines ausländischen Vertreters untersagen will, obwohl der Parkplatz groß genug und die Polizei ausreichend gegen Ausschreitungen gewappnet ist? ...
Nachruf auf John Neubauer

Am 5. Oktober 2015 ist in Amsterdam der Germanist und Komparatist John Neubauer verstorben, an einer erst kurz zuvor diagnostizierten und tapfer ertragenen schweren Krankheit, im Kreise seiner Angehörigen und im Alter von nicht ganz zweiundachtzig Jahren. Unser Fach verliert in ihm einen hervorragenden Vertreter, und alle, die ihn kannten, verlieren einen hochsympathischen und inspirierenden Mitmenschen.
Der vorliegende Beitrag beschäftigt sich mit einem Gedicht Heiner Müllers, das dieser in unmittelbarer Reaktion auf den Zusammenbruch des osteuropäischen Sozialismus verfasste. Da dieses Gedicht in mehreren Etappen geschrieben und publiziert wurde, gibt es Einblicke in die von Umbruch und Verwerfungen gekennzeichnete Entstehungssituation, die der Beitrag erläutern will.
Es kann gleichermaßen als eine vorläufige Lebensbilanz des Autors gelesen werden, wo sich selbstkritische Äußerungen zu den Irrtümern mit denen einer geschichtlichen Epoche kreuzen. Da die eingenommene Haltung zu diesem komplexen Veränderungsprozess und die Bewertung dieser Wende durch den Autor auch wiederum einer Selbstkritik unterworfen wurde, versucht der Beitrag, einen Einblick in die geschichtsphilosophische und ästhetische Werkstatt des Autors Heiner Müller zu geben
Treatment of refractory ascites with an automated low-flow ascites pump in patients with cirrhosis

Background: Refractory ascites (RA) is a frequent complication of cirrhosis, requiring large volume paracentesis or placement of a transjugular intrahepatic portosystemic shunt (TIPSS). The automated low-flow ascites pump (alfapump, Sequana Medical AG, Zurich, Switzerland) is an innovative treatment option for patients with RA.
Aim: To assess safety and efficacy of this treatment in patients with a contraindication to TIPSS.
Methods: Fifty-six patients (43 males; mean age 62 years) from centres in Germany, Switzerland, UK and Spain were included and followed for up to 24 months. Complications, device deficiencies, paracentesis frequency and patient survival were recorded.
Results: At the time of this analysis, 3 patients completed the 24-month observation period, monitoring of 3 was ongoing, 9 underwent liver transplantation, 17 patients were withdrawn due to serious adverse events and 23 patients died. Most frequently observed technical complication was blocking of the peritoneal catheter. Twenty-three pump-related reinterventions (17 patients) and 12 pump exchanges (11 patients) were required during follow-up. The pump system was explanted in 48% of patients (in 17 patients due to serious adverse events, in 9 at the time of liver transplantation and in 1 due to recovery from RA). Median frequency of paracentesis dropped from 2.17 to 0.17 per month.
Conclusions: The alfapump can expand therapeutic options for cirrhotic patients with RA. Continuous drainage of ascites in a closed loop automated system led to significant reduction in paracentesis frequency. Technical and procedural improvements are required to reduce the rate of adverse events and reinterventions.
Das Recht – abstrakt und bilderfeindlich? Ein Fehlurteil. Die heutige Bilderflut ist jedoch auch für die Rechtswissenschaft ein neues Phänomen.
Anticoagulation with warfarin and rivaroxaban ameliorates experimental autoimmune encephalomyelitis

Background: In multiple sclerosis, coagulation factors have been shown to modulate inflammation. In this translational study, we investigated whether long-term anticoagulation with warfarin or rivaroxaban has beneficial effects on the course of autoimmune experimental encephalomyelitis (EAE).
Methods: Female SJL/J mice treated with anticoagulants namely warfarin or rivaroxaban were immunized with PLP139–151. Stable anticoagulation was maintained throughout the entire experiment. Mice without anticoagulation treated with the vehicle only were used as controls. The neurological deficit was recorded during the course of EAE, and histopathological analyses of inflammatory lesions were performed.
Results: In preventive settings, both treatment with warfarin and rivaroxaban reduced the maximum EAE score as compared to the control group and led to a reduction of inflammatory lesions in the spinal cord. In contrast, therapeutic treatment with warfarin had no beneficial effects on the clinical course of EAE. Signs of intraparenchymal hemorrhage at the site of the inflammatory lesions were not observed.
Conclusion: We developed long-term anticoagulation models that allowed exploring the course of EAE under warfarin and rivaroxaban treatment. We found a mild preventive effect of both warfarin and rivaroxaban on neurological deficits and local inflammation, indicating a modulation of the disease induction by anticoagulation.
Nutrition support is a necessary therapy for critically ill cardiac surgery patients. However, conclusive evidence for this population, consisting of well-conducted clinical trials is lacking. To clarify optimal strategies to improve outcomes, an international multidisciplinary group of 25 experts from different clinical specialties from Germany, Canada, Greece, USA and Russia discussed potential approaches to identify patients who may benefit from nutrition support, when best to initiate nutrition support, and the potential use of pharmaco-nutrition to modulate the inflammatory response to cardiopulmonary bypass. Despite conspicuous knowledge and evidence gaps, a rational nutritional support therapy is presented to benefit patients undergoing cardiac surgery.
Gene trap mutagenesis is a powerful tool to create loss-of-function mutations in mice and other model organisms. Modifications of traditional gene trap cassettes, including addition of conditional features in the form of Flip-excision (FlEx) arrays to enable directional gene trap cassette inversions by Cre and Flpe site-specific recombinases, greatly enhanced their experimental potential. By taking advantage of these conditional gene trap cassettes, we developed a generic strategy for generating conditional mutations and validated this strategy in mice carrying a multipurpose allele of the Prdm16 transcription factor gene. We demonstrate that the gene trap insertion creates a null mutation replicating the Pierre Robin sequence-type cleft palate phenotype of other Prdm16 mutant mice. Consecutive breeding to Flpe and Emx1IREScre deleter mice spatially restricted Prdm16 loss to regions of the forebrain expressing the homeobox gene Emx1, demonstrating the utility of the technology for the analysis of tissue-specific gene functions.
Background: In oldest-old patients (>80), few trials showed efficacy of treating hypertension and they included mostly the healthiest elderly. The resulting lack of knowledge has led to inconsistent guidelines, mainly based on systolic blood pressure (SBP), cardiovascular disease (CVD) but not on frailty despite the high prevalence in oldest-old. This may lead to variation how General Practitioners (GPs) treat hypertension. Our aim was to investigate treatment variation of GPs in oldest-olds across countries and to identify the role of frailty in that decision.
Methods: Using a survey, we compared treatment decisions in cases of oldest-old varying in SBP, CVD, and frailty. GPs were asked if they would start antihypertensive treatment in each case. In 2016, we invited GPs in Europe, Brazil, Israel, and New Zealand. We compared the percentage of cases that would be treated per countries. A logistic mixed-effects model was used to derive odds ratio (OR) for frailty with 95% confidence intervals (CI), adjusted for SBP, CVD, and GP characteristics (sex, location and prevalence of oldest-old per GP office, and years of experience). The mixed-effects model was used to account for the multiple assessments per GP.
Results: The 29 countries yielded 2543 participating GPs: 52% were female, 51% located in a city, 71% reported a high prevalence of oldest-old in their offices, 38% and had >20 years of experience. Across countries, considerable variation was found in the decision to start antihypertensive treatment in the oldest-old ranging from 34 to 88%. In 24/29 (83%) countries, frailty was associated with GPs’ decision not to start treatment even after adjustment for SBP, CVD, and GP characteristics (OR 0.53, 95%CI 0.48–0.59; ORs per country 0.11–1.78).
Conclusions: Across countries, we found considerable variation in starting antihypertensive medication in oldest-old. The frail oldest-old had an odds ratio of 0.53 of receiving antihypertensive treatment. Future hypertension trials should also include frail patients to acquire evidence on the efficacy of antihypertensive treatment in oldest-old patients with frailty, with the aim to get evidence-based data for clinical decision-making.
Non-lethal genotyping of Tribolium castaneum adults using genomic DNA extracted from wing tissue

The red flour beetle Tribolium castaneum has become the second most important insect model organism and is frequently used in developmental biology, genetics and pest-associated research. Consequently, the methodological arsenal increases continuously, but many routinely applied techniques for Drosophila melanogaster and other insect species are still unavailable. For example, a protocol for non-lethal genotyping has not yet been adapted but is particularly useful when individuals with known genotypes are required for downstream experiments. In this study, we present a workflow for non-lethal genotyping of T. castaneum adults based on extracting genomic DNA from wing tissue. In detail, we describe a convenient procedure for wing dissection and a custom method for wing digestion that allows PCR-based genotyping of up to fifty adults in less than an afternoon with a success rate of about 86%. The amount of template is sufficient for up to ten reactions while viability and fertility of the beetles are preserved. We prove the applicability of our protocol by genotyping the white / scarlet gene pair alleles from the black-eyed San Bernadino wild-type and white-eyed Pearl recessive mutant strains spanning four generations. Non-lethal genotyping has the potential to improve and accelerate many workflows: Firstly, during the establishment process of homozygous cultures or during stock keeping of cultures that carry recessively lethal alleles, laborious test crossing is replaced by non-lethal genotyping. Secondly, in genome engineering assays, non-lethal genotyping allows the identification of appropriate founders before they are crossed against wild-types, narrowing the efforts down to only the relevant individuals. Thirdly, non-lethal genotyping simplifies experimental strategies, in which genotype and behavior should be correlated, since the genetic configuration of potential individuals can be determined before the actual behavior assays is performed.
Purpose: To evaluate long-term outcome of three years and treatment patterns of patients suffering from severely drug-refractory epilepsy (SDRE).
Methods: This analysis was population-based and retrospective, with data collected from four million individuals insured by statutory German health insurance. ICD-10 codes for epilepsy (G40*) and intake of anticonvulsants were used to identify prevalent cases, which were then compared with a matched cohort drawn from the population at large. Insurance data were available from 2008 to 2013. Any patient who had been prescribed with at least four different antiepileptic drugs (AEDs) in an 18-month period was defined as an SDRE case.
Results: A total of 769 patients with SDRE were identified. Of these, 19% were children and adolescents; the overall mean age was 42.3 years, 45.4% were female and 54.6% male. An average of 2.7 AEDs per patient was prescribed during the first follow-up year. The AEDs most commonly prescribed were: levetiracetam (53.5%), lamotrigine (41.4%), valproate (41.3%), lacosamide (20.4%), and topiramate (17.8%). During 3-year follow-up, there was an annual rate of hospitalization in the range 42.7 to 55%, which was significantly higher than the 11.6–12.8% (p < 0.001) for the matched controls. Admissions to hospital because of epilepsy ranged between 1.7 and 1.9 per year, with an average duration for each epilepsy-caused hospitalization of 10–11.1 days. The number of comorbidities for SDRE patients was significantly increased compared with the matched controls: depression (28% against 10%), vascular disorders (22% against 5%), and injury rates were also higher (head 16% against 3%, trunk and limbs 16% against 8%). The 3-year mortality rate for SDRE patients was 14% against 2.1% in the matched cohort.
Conclusion: SDRE patients are treated with AED polytherapy for all of the 3-year follow-up period. They are hospitalized more frequently than the general population and show increased morbidity levels and a sevenfold increase in mortality rate over 3 years. Further examination is required of ways in which new approaches to treatment could lead to better outcomes in severely affected patients.
Background: Recognizing patients at risk for pulmonary complications (PC) is of high clinical relevance. Migration of polymorphonuclear leukocytes (PMN) to inflammatory sites plays an important role in PC, and is tightly regulated by specific chemokines including interleukin (IL)−8 and other mediators such as leukotriene (LT)B4. Previously, we have reported that LTB4 indicated early patients at risk for PC after trauma. Here, the relevance of LTB4 to indicating lung integrity in a newly established long-term porcine severe trauma model (polytrauma, PT) was explored.
Methods: mTwelve pigs (3 months old, 30 ± 5 kg) underwent PT including standardized femur fracture, lung contusion, liver laceration, hemorrhagic shock, subsequent resuscitation and surgical fracture fixation. Six animals served as controls (sham). After 72 h lung damage and inflammatory changes were assessed. LTB4 was determined in plasma before the experiment, immediately after trauma, and after 2, 4, 24 or 72 h. Bronchoalveolar lavage (BAL)-fluid was collected prior and after the experiment.
Results: Lung injury, local gene expression of IL-8, IL-1β, IL-10, IL-18 and PMN-infiltration into lungs increased significantly in PT compared with sham. Systemic LTB4 increased markedly in both groups 4 h after trauma. Compared with declined plasma LTB4 levels in sham, LTB4 increased further in PT after 72 h. Similar increase was observed in BAL-fluid after PT.
Conclusions: In a severe trauma model, sustained changes in terms of lung injury and inflammation are determined at day 3 post-trauma. Specifically, increased LTB4 in this porcine long-term model indicated a rapid inflammatory alteration both locally and systemically. The results support the concept of LTB4 as a biomarker for PC after severe trauma and lung contusion.
Der Begriff 'apokryph' taucht bei Johann Georg Hamann (1730-1788), dem 'Magus in Norden', seltener auf als man es vielleicht erwarten würde, jedenfalls so man Hamann noch immer nach dem gängigen Klischee für 'dunkel', einen 'Mystiker' oder einen 'Obskurantisten' oder gar für den Ursprung des modernen und spezifisch deutschen Irrationalismus und extremsten Feind der Aufklärung hält, gegenüber dessen "gewollter und künstlicher Dunkelheit", die er wie eine "Nebelwand um sich zog", sogar Hegels Phänomenologie des Geistes "eine wahre Ferienlektüre" sei. Ein schöpferischer Kopf also, "der manche tiefsinnigen Einfälle hatte, aber in welch geheimnisvolle Gewänder pflegte er sie zu verkleiden!" Entsprechend ist Hamanns Ruf: "Dunkelheit ist das traditionelle Prädicat, das jedem deutschen Primaner über ihn geläufig ist." Mit seiner Dunkelheit, so die unausgesprochene Annahme, möchte Hamann willentlich und wissentlich etwas verbergen, vielleicht nicht nur bei sich, sondern letztlich sogar das Licht der Aufklärung überhaupt; ein Umstand, der auf das Schönste zum griechischen apokryptein: 'verbergen, verdunkeln' (so die Übersetzung in Josef Nadlers Schlüssel zu Hamanns Werken) zu passen scheint. Entsprechend ist es, um eine weitere Bedeutung des Begriffes "apokryph" als des 'Gegen-Kanonischen' heranzuziehen, kaum verwunderlich, dass Texte Hamanns es nicht in den akademisch etablierten Kanon philosophischer Werke geschafft haben, ja sogar gegen entsprechende kanonische Texte (etwa die Metakritik über den Purismum der Vernunft gegen die Kritik der reinen Vernunft oder Golgatha und Scheblimini gegen Mendelssohns Jerusalem) gerichtet zu sein scheinen.
I summarize recent developments in the hard-thermal-loop approach to QCD. I first discuss a finite-temperature and -density calculation of QCD thermodynamics at NNLO from the hard-thermal-loop perturbation theory. I then discuss a generalization of the hard-thermal-loop framework to the magnetic scale g2T, from which a novel non-Abelian massless mode is uncovered.
Evidence of widespread wildfires in a coal seam from the middle Permian of the North China Basin

The North China Basin is the largest coal-bearing basin in China, and has an areal extent of 800,000 km2. We analyzed 138 coal samples and in situ pillar coal samples of the middle Permian from this basin by macropetrography, microscope, scanning electron microscope, gas chromatography, and gas chromatography–mass spectrometer in order to study wildfires. High contents of inertinite (charcoal) and natural coke particles observed in coal samples indicate that vegetation in precursor mires and peats of the middle Permian coal from north China was exposed to far-ranging wildfires. In addition, high-molecular-weight polycyclic aromatic hydrocarbons were detected in the coal samples. These aromatic compounds were formed under high temperatures and provide further evidence of wildfire. These wildfires would have discharged significant CO and CO2 gases into the atmosphere and affected the paleoclimate and paleoecosystem.
Background: The Ebola virus has been responsible for numerous outbreaks since the 1970s, with the most recent outbreak taking place between 2014 and 2016 and causing an international public health emergency. Ebola virus disease (EVD) has a high mortality rate and no approved targeted treatment exists to date. A number of established drugs are being considered as potential therapeutic agents for the treatment of EVD.
Objective: We aimed to identify potential drug repositioning candidates and to assess the scientific evidence available on their efficacy.
Methods: We conducted a systematic literature search in MEDLINE, Embase, and other relevant trial registry platforms for studies published between January 1976 and January 2017. We included drug screening, preclinical studies, and clinical studies on repurposed drugs for the treatment of EVD. The risk of bias for animal studies and nonrandomized clinical studies was assessed. The quality of reporting for case series and case reports was evaluated. Finally, we selected drugs approved by established regulatory authorities, which have positive in vitro study outcomes and at least one additional animal or clinical trial.
Results: We identified 3301 publications, of which 37 studies fulfilled our inclusion criteria. Studies were highly heterogeneous in terms of study type, methodology, and intervention. The risk of bias was high for 13 out of 14 animal studies. We selected 11 drugs with potential anti-EVD therapeutic effects and summarized their evidence.
Conclusions: Several established drugs may have therapeutic effects on EVD, but the quality and quantity of current scientific evidence is lacking. This review highlights the need for well-designed and conducted preclinical and clinical research to establish the efficacy of potential repurposed drugs against EVD.
S1P provokes tumor lymphangiogenesis via macrophage-derived mediators such as IL-1β or lipocalin-2

A pleiotropic signaling lipid, sphingosine-1-phosphate (S1P), has been implicated in various pathophysiological processes supporting tumor growth and metastasis. However, there are only a few descriptive studies suggesting a role of S1P in tumor lymphangiogenesis, which is critical for tumor growth and dissemination. Corroborating own data, the literature suggests that apoptotic tumor cell-derived S1P alters the phenotype of tumor-associated macrophages (TAMs) to gain protumor functions. However, mechanistically, the role of TAM-induced lymphangiogenesis has only been poorly described, mostly linked to the production of lymphangiogenic factors such as vascular endothelial growth factor C (VEGF-C) and VEGF-D, or transdifferentiation into lymphatic endothelial cells. Recent findings highlight a rather underappreciated role of S1P in tumor lymphangiogenesis, referring to the production of interleukin-1β (IL-1β) and lipocalin-2 (LCN2) by a tumor-promoting macrophage phenotype. In this review, we aim to provide to the readers with the current understanding of the molecular mechanism how apoptotic cell-derived S1P triggers TAMs to promote lymphangiogenesis.
The aim of our study was to reveal the differences in the species composition and soil properties of open sandy grasslands dominated by different Festuca species, Festuca vaginata and F. pseudovaginata. Due to the arid conditions, sandy grasslands are generally covered by xerothermic vegetation in which F. vaginata is a typical dominant species. Festuca pseudovaginata, a species newly described by the authors, can also gain dominance in sandy grasslands. However, species composition and soil properties of grasslands dominated by this recently discovered species are still undiscovered. Based on previous coenological studies, we hypothesised that the grasslands characterised by the endemic F. vaginata are more species-rich than those with F.pseudovaginata.
Coenological sampling was carried out in May 2009 at two study sites using the Braun-Blanquet method in quadrats of 2 m × 2 m. Five relevés were sampled in stands dominated by F. vaginata and F. pseudovaginata respectively at two study sites in Central Hungary, resulting in a total of twenty relevés. Analyses were based on the cover scores of vascular plant species and cryptogam crust and values of seven soil properties (pH [KCl], pH [H2O], humus, total N, Ca, P2O5 and K2O) measured in the 0–15 and 15–30 cm soil layers. Soil properties of the grasslands dominated by F. pseudovaginata and F. vaginata (dependent variables) were compared by linear mixed models, where ‘grassland type’ was the fixed factor and ‘site nested in grassland type’ was considered as random factor. Data were analysed by cluster analysis, fusion algorithm was a combinatorial method (minimising increase of variance), and the correlation was used as comparative function. We compared the cover of subordinate species by Mann-Whitney U test. We found that F. pseudovaginata and F. vaginata samples were well separated, and grasslands dominated by F. pseudovaginata had nearly two times more species than those dominated by F. vaginata. Based on the cluster analyses using plant cover and upper 0–15 cm soil layer data, F. pseudovaginata and F. vaginata groups could be well separated. Linear mixed models revealed that F. vaginata grasslands were typical on soils with higher pH, nitrogen, phosphorous and potassium contents compared to F. pseudovaginata grasslands, which indicates a tight connection between the properties of the upper soil layer and the vegetation in sandy grasslands.
Dort, wo es zu einem intensiven kulturellen Kontakt, Austausch und Transfer zwischen Gemeinschaften kommt, lassen sich Kategorien wie Identität und Alterität nicht immer scharf voneinander trennen, sondern erscheinen eher als "ständig zu aktualisierende soziale Konstruktionen" (NEULAND 2013: 168). Sie können zudem auch zu transkulturellen innovativen sprachlichen Schöpfungen führen. Die Sprache ist wegen ihrer identitätskonstitutiven Funktion fester Bestandteil des Modells der multiplen Sprachidentität von Marijana KRESIĆ (2007). Ausgehend von diesem Modell wird im Beitrag das enge Verhältnis von Identitätskonstruktion - Standardsprache - Sprachgebrauch diskutiert und anhand von ausgewählten Beispielen dargestellt. Die identitätskonstitutive Funktion der Sprache basiert auf der Selektion von Normen aus dem Sprachsystem, wobei der Sprechende durch die Verwendung sprachlicher Zeichen, "seine (soziale und/oder personale Identität) [begründet], d.h. er markiert, wer er als Individuum ist bzw. welcher sozialen Gruppe er zugehört" (KRESIĆ 2007: 19). Im Rahmen der Besprechung dieses Konzepts wird geprüft, wie Formen interkulturellen Sprachgebrauchs (z. B. Kiezdeutsch) in dieses Modell eingefügt werden können.
Broad AOX expression in a genetically tractable mouse model does not disturb normal physiology

Plants and many lower organisms, but not mammals, express alternative oxidases (AOXs) that branch the mitochondrial respiratory chain, transferring electrons directly from ubiquinol to oxygen without proton pumping. Thus, they maintain electron flow under conditions when the classical respiratory chain is impaired, limiting excess production of oxygen radicals and supporting redox and metabolic homeostasis. AOX from Ciona intestinalis has been used to study and mitigate mitochondrial impairments in mammalian cell lines, Drosophila disease models and, most recently, in the mouse, where multiple lentivector-AOX transgenes conferred substantial expression in specific tissues. Here, we describe a genetically tractable mouse model in which Ciona AOX has been targeted to the Rosa26 locus for ubiquitous expression. The AOXRosa26 mouse exhibited only subtle phenotypic effects on respiratory complex formation, oxygen consumption or the global metabolome, and showed an essentially normal physiology. AOX conferred robust resistance to inhibitors of the respiratory chain in organello; moreover, animals exposed to a systemically applied LD50 dose of cyanide did not succumb. The AOXRosa26 mouse is a useful tool to investigate respiratory control mechanisms and to decipher mitochondrial disease aetiology in vivo.
Robinia pseudoacacia L. (black locust) is a North American tree, considered controversial because of the conflict between multiple uses by humans and negative environmental impacts, which have resulted in it being listed among the most invasive species in Europe. The current management of Robinia stands in Central Europe varies locally according to national legislation, preferring either socio-economic benefits or biodiversity impacts. We collected field data from our target region of Czechia, reviewed research articles including local grey literature mostly from Central and Southern Europe, unpublished results of local projects and inquired relevant specialists. Because Robinia grows in habitats ranging from urban to forest to natural grassland, neither unrestricted cultivation nor large-scale eradication is applicable as a universal practice. In this paper we suggest a complex management strategy for Robinia stands that takes into account habitat, this species’ local ability to spread, as well as economic, cultural and biodiversity aspects. We categorized Robinia stands growing in Europe into eight groups and proposed stratified approach to the management based on decisions that reflect local context. Depending on that, the management includes (i) establishment of new plantations, (ii) maintenance or utilization of existing stands, (iii) tolerance
and (iv) conversion to original vegetation. Our complex management strategy will provide a comprehensive guideline for the management of alien trees in Europe.

The CRISPR/Cas9 prokaryotic adaptive immune system and its swift repurposing for genome editing enables modification of any prespecified genomic sequence with unprecedented accuracy and efficiency, including targeted gene repair. We used the CRISPR/Cas9 system for targeted repair of patient-specific point mutations in the Cytochrome b-245 heavy chain gene (CYBB), whose inactivation causes chronic granulomatous disease (XCGD)—a life-threatening immunodeficiency disorder characterized by the inability of neutrophils and macrophages to produce microbicidal reactive oxygen species (ROS). We show that frameshift mutations can be effectively repaired in hematopoietic cells by non-integrating lentiviral vectors carrying RNA-guided Cas9 endonucleases (RGNs). Because about 25% of most inherited blood disorders are caused by frameshift mutations, our results suggest that up to a quarter of all patients suffering from monogenic blood disorders could benefit from gene therapy employing personalized, donor template-free RGNs.
It is well known that the media has strongly shaped the life of the people in many respects and that it exerts a sustained influence on our value systems and ways of thinking. Thus it also shows a clear extension of the human life. By far less famously however is the fact that nowadays media is the basis for all forms of mental development. This is why the relation between the people and the media is extremely tightened. Nevertheless, this ambivalent relation offers multi-complex material for the literary inspiration. Addressing the topic of the media in the literature combines two aspects ritically: On the one hand it reflects the human behaviour compared with the media and, on the other hand, it underlines emphatically the intermedial writing itself. My speech will be dedicated to the question: how these both aspects interact with each other. The narrative text of Alfred Andersch’s Erinnerte Gestalten will serve the answers to my topic. In this three prosaic texts Andersch shows the subject of the intermedial writing from different perspectives and he discusses certain human reactions to the media.
Background: To meet the requirements imposed by the time-dependency of acute stroke therapies, it is necessary 1) to initiate structural and cultural changes in the breadth of stroke-ready hospitals and 2) to find new ways to train the personnel treating patients with acute stroke. We aimed to implement and validate a composite intervention of a stroke team algorithm and simulation-based stroke team training as an effective quality initiative in our regional interdisciplinary neurovascular network consisting of 7 stroke units.
Methods: We recorded door-to-needle times of all consecutive stroke patients receiving thrombolysis at seven stroke units for 3 months before and after a 2 month intervention which included setting up a team-based stroke workflow at each stroke unit, a train-the-trainer seminar for stroke team simulation training and a stroke team simulation training session at each hospital as well as a recommendation to take up regular stroke team trainings.
Results: The intervention reduced the network-wide median door-to-needle time by 12 minutes from 43,0 (IQR 29,8–60,0, n = 122) to 31,0 (IQR 24,0–42,0, n = 112) minutes (p < 0.001) and substantially increased the share of patients receiving thrombolysis within 30 minutes of hospital arrival from 41.5% to 59.6% (p < 0.001). Stroke team training participants stated a significant increase in knowledge on the topic of acute stroke care and in the perception of patient safety. The overall course concept was regarded as highly useful by most participants from different professional backgrounds.
Conclusions: The composite intervention of a binding team-based algorithm and stroke team simulation training showed to be well-transferable in our regional stroke network. We provide suggestions and materials for similar campaigns in other stroke networks.
Background: The Hawking–Perry–Strominger (HPS) work states a new controversial idea about the black hole (BH) information paradox , where BHs maximally entropize and encode information in their event horizon area , with no “hair” thought to reveal information outside but angular momentum, mass, and electric charge only in a unique quantum gravity (QG) vacuum state. New conservation laws of gravitation and electromagnetism , appear to generate different QG vacua, preserving more information in soft photon/graviton hair implants. We find that BH photon hair implants can encode orbital angular momentum (OAM) and vorticity of the electromagnetic (EM) field. Methods: Numerical simulations are used to plot an EM field with OAM emitted by a set of dipolar currents together with the soft photon field they induce. The analytical results confirm that the soft photon hair implant carries OAM and vorticity. Results: a set of charges and currents generating real EM fields with precise values of OAM induce a “curly”, twisted, soft-hair implant on the BH with vorticity and OAM increased by one unit with respect to the initial real field. Conclusions: Soft photon implants can be spatially shaped ad hoc, encoding structured and densely organized information on the event horizon
Background: Root and tuber crops are a major food source in tropical Africa. Among these crops are several species in the monocotyledonous genus Dioscorea collectively known as yam, a staple tuber crop that contributes enormously to the subsistence and socio-cultural lives of millions of people, principally in West and Central Africa. Yam cultivation is constrained by several factors, and yam can be considered a neglected “orphan” crop that would benefit from crop improvement efforts. However, the lack of genetic and genomic tools has impeded the improvement of this staple crop.
Results: To accelerate marker-assisted breeding of yam, we performed genome analysis of white Guinea yam (Dioscorea rotundata) and assembled a 594-Mb genome, 76.4% of which was distributed among 21 linkage groups. In total, we predicted 26,198 genes. Phylogenetic analyses with 2381 conserved genes revealed that Dioscorea is a unique lineage of monocotyledons distinct from the Poales (rice), Arecales (palm), and Zingiberales (banana). The entire Dioscorea genus is characterized by the occurrence of separate male and female plants (dioecy), a feature that has limited efficient yam breeding. To infer the genetics of sex determination, we performed whole-genome resequencing of bulked segregants (quantitative trait locus sequencing [QTL-seq]) in F1 progeny segregating for male and female plants and identified a genomic region associated with female heterogametic (male = ZZ, female = ZW) sex determination. We further delineated the W locus and used it to develop a molecular marker for sex identification of Guinea yam plants at the seedling stage.
Conclusions: Guinea yam belongs to a unique and highly differentiated clade of monocotyledons. The genome analyses and sex-linked marker development performed in this study should greatly accelerate marker-assisted breeding of Guinea yam. In addition, our QTL-seq approach can be utilized in genetic studies of other outcrossing crops and organisms with highly heterozygous genomes. Genomic analysis of orphan crops such as yam promotes efforts to improve food security and the sustainability of tropical agriculture.
Thirty-seven alien plant species, pre-identified by horizon scanning exercises were prioritised for pest risk analysis (PRA) using a modified version of the EPPO Prioritisation Process designed to be compliant with the EU Regulation 1143/2014. In Stage 1, species were categorised into one of four lists – a Residual List, EU List of Minor Concern, EU Observation List and the EU List of Invasive Alien Plants. Only those species included in the latter proceeded to the risk management stage where their priority for PRA was assessed. Due to medium or high spread potential coupled with high impacts twenty-two species were included in the EU List of Invasive Alien Plants and proceeded to Stage 2. Four species (Ambrosia trifida, Egeria densa, Fallopia baldschuanica and Oxalis pes-caprae) were assigned to the EU Observation List due to moderate or low impacts. Albizia lebbeck, Clematis terniflora, Euonymus japonicus, Lonicera morrowii, Prunus campanulata and Rubus rosifolius were assigned to the residual list due to a current lack of information on impacts. Similarly, Cornus sericea and Hydrilla verticillata were assigned to the Residual List due to unclear taxonomy and uncertainty in native status, respectively. Chromolaena odorata, Cryptostegia grandiflora and Sphagneticola trilobata were assigned to the Residual List as it is unlikely they will establish in the Union under current climatic conditions. In the risk management stage, Euonymus fortunei, Ligustrum sinense and Lonicera maackii were considered a low priority for PRA as they do not exhibit invasive tendencies despite being widely cultivated in the EU over several decades. Nineteen species were identified as having a high priority for a PRA (Acacia dealbata, Ambrosia confertiflora, Andropogon virginicus, Cardiospermum grandiflorum, Celastrus orbiculatus, Cinnamomum camphora, Cortaderia jubata, Ehrharta calycina, Gymnocoronis spilanthoides, Hakea sericea, Humulus scandens, Hygrophila polysperma, Lespedeza cuneata, Lygodium japonicum, Pennisetum setaceum, Prosopis juliflora, Sapium sebiferum, Pistia stratiotes and Salvinia molesta).
Carbon mass of the non-indigenous predatory fishhook water flea Cercopagis pengoi (Ostroumov, 1891) from the eastern Gulf of Finland, the Baltic Sea, was for the first time measured using the high temperature combustion method. Prior to the analysis, individual dry weight of Cercopagis was determined; altogether ca. 500 organisms were examined. Mean individual dry weight of C. pengoi for July-September was estimated as 34.0 μg; carbon mass averaged 15.8 μg; carbon content, calculated as percent of dry weight, averaged 43.4%. Those values varied over months, mainly because of different population structure of C. pengoi and variation in their diet due to seasonal dynamics of the food objects. However, relations between carbon mass and dry weight for different months did not differ statistically (p<0.001). Therefore, the general polynomial regressions (k=2), describing carbon mass-to-dry weight and carbon content-to-dry weight relationships, were calculated for the entire dataset of individual measurements of C. pengoi body metrics. These data will contribute to adequate evaluation of food web structure and ecosystem alterations in various water bodies invaded by C. pengoi which has got a strong potential to pelagic food web transformations that may impact the overall energy balance and decrease the size of fish stocks.
Natural sounds convey perceptually relevant information over multiple timescales, and the necessary extraction of multi-timescale information requires the auditory system to work over distinct ranges. The simplest hypothesis suggests that temporal modulations are encoded in an equivalent manner within a reasonable intermediate range. We show that the human auditory system selectively and preferentially tracks acoustic dynamics concurrently at 2 timescales corresponding to the neurophysiological theta band (4–7 Hz) and gamma band ranges (31–45 Hz) but, contrary to expectation, not at the timescale corresponding to alpha (8–12 Hz), which has also been found to be related to auditory perception. Listeners heard synthetic acoustic stimuli with temporally modulated structures at 3 timescales (approximately 190-, approximately 100-, and approximately 30-ms modulation periods) and identified the stimuli while undergoing magnetoencephalography recording. There was strong intertrial phase coherence in the theta band for stimuli of all modulation rates and in the gamma band for stimuli with corresponding modulation rates. The alpha band did not respond in a similar manner. Classification analyses also revealed that oscillatory phase reliably tracked temporal dynamics but not equivalently across rates. Finally, mutual information analyses quantifying the relation between phase and cochlear-scaled correlations also showed preferential processing in 2 distinct regimes, with the alpha range again yielding different patterns. The results support the hypothesis that the human auditory system employs (at least) a 2-timescale processing mode, in which lower and higher perceptual sampling scales are segregated by an intermediate temporal regime in the alpha band that likely reflects different underlying computations.
Peer review of research articles is a core part of our scholarly communication system. In spite of its importance, the status and purpose of peer review is often contested. What is its role in our modern digital research and communications infrastructure? Does it perform to the high standards with which it is generally regarded? Studies of peer review have shown that it is prone to bias and abuse in numerous dimensions, frequently unreliable, and can fail to detect even fraudulent research. With the advent of Web technologies, we are now witnessing a phase of innovation and experimentation in our approaches to peer review. These developments prompted us to examine emerging models of peer review from a range of disciplines and venues, and to ask how they might address some of the issues with our current systems of peer review. We examine the functionality of a range of social Web platforms, and compare these with the traits underlying a viable peer review system: quality control, quantified performance metrics as engagement incentives, and certification and reputation. Ideally, any new systems will demonstrate that they out-perform current models while avoiding as many of the biases of existing systems as possible. We conclude that there is considerable scope for new peer review initiatives to be developed, each with their own potential issues and advantages. We also propose a novel hybrid platform model that, at least partially, resolves many of the technical and social issues associated with peer review, and can potentially disrupt the entire scholarly communication system. Success for any such development relies on reaching a critical threshold of research community engagement with both the process and the platform, and therefore cannot be achieved without a significant change of incentives in research environments.

Drug induced liver injury (DILI) is a potentially serious adverse reaction in a few susceptible individuals under therapy by various drugs. Health care professionals facing DILI are confronted with a wealth of drug-unrelated liver diseases with high incidence and prevalence rates, which can confound the DILI diagnosis. Searching for alternative causes is a key element of RUCAM (Roussel Uclaf Causality Assessment Method) to assess rigorously causality in suspected DILI cases. Diagnostic biomarkers as blood tests would be a great help to clinicians, regulators, and pharmaceutical industry would be more comfortable if, in addition to RUCAM, causality of DILI can be confirmed. High specificity and sensitivity are required for any diagnostic biomarker. Although some risk factors are available to evaluate liver safety of drugs in patients, no valid diagnostic or prognostic biomarker exists currently for idiosyncratic DILI when a liver injury occurred. Identifying a biomarker in idiosyncratic DILI requires detailed knowledge of cellular and biochemical disturbances leading to apoptosis or cell necrosis and causing leakage of specific products in blood. As idiosyncratic DILI is typically a human disease and hardly reproducible in animals, pathogenetic events and resulting possible biomarkers remain largely undisclosed. Potential new diagnostic biomarkers should be evaluated in patients with DILI and RUCAM-based established causality. In conclusion, causality assessment in cases of suspected idiosyncratic DILI is still best achieved using RUCAM since specific biomarkers as diagnostic blood tests that could enhance RUCAM results are not yet available.
AKTIVA-MCI is a program for patients with mild cognitive impairment (MCI) that aims to enhance participation in cognitively stimulating leisure activities. Participation in cognitively stimulating activities seems to be a potential strategy for people with MCI delaying cognitive decline for a while. In total, 35 MCI patients were enrolled in the pilot study of whom 29 completed the whole program (16 female, 71.1±7.5 years; Mini Mental Status Examination score: 28±2.2). Daily activity protocols were used to measure the frequency of participation in cognitively stimulating activities during the program (12 sessions). Additional standardized psychometric tests and questionnaires were used to assess cognition, mood, and subjective memory decline. Analyses of the daily activity protocols showed that during the intervention participants increased the frequency of several cognitively stimulating leisure activities. Comparison of pre-post data indicates no changes in cognitive status, mood, and subjective memory decline. These findings indicate that the program is suitable for patients with MCI.
The adaptor protein Src homology 2 domain-containing leukocyte phosphoprotein of 76 kDa (SLP-76) plays a crucial role in T cell activation by linking antigen receptor (T cell receptor, TCR) signals to downstream pathways. At its N terminus, SLP-76 has three key tyrosines (Tyr-113, Tyr-128, and Tyr-145, “3Y”) as well as a sterile α motif (SAM) domain whose function is unclear. We showed previously that the SAM domain has two binding regions that mediate dimer and oligomer formation. In this study, we have identified SAM domain-carrying non-receptor tyrosine kinase, activated Cdc42-associated tyrosine kinase 1 (ACK1; also known as Tnk2, tyrosine kinase non-receptor 2) as a novel binding partner of SLP-76. Co-precipitation, laser-scanning confocal microscopy, and in situ proximity analysis confirmed the binding of ACK1 to SLP-76. Further, the interaction was induced in response to the anti-TCR ligation and abrogated by the deletion of SLP-76 SAM domain (ΔSAM) or mutation of Tyr-113, Tyr-128, and Tyr-145 to phenylalanine (3Y3F). ACK1 induced phosphorylation of the SLP-76 N-terminal tyrosines (3Y) dependent on the SAM domain. Further, ACK1 promoted calcium flux and NFAT-AP1 promoter activity and decreased the motility of murine CD4+ primary T cells on ICAM-1-coated plates, an event reversed by a small molecule inhibitor of ACK1 (AIM-100). These findings identify ACK1 as a novel SLP-76-associated protein-tyrosine kinase that modulates early activation events in T cells.

The field of dynamic nuclear polarization has undergone tremendous developments and diversification since its inception more than 6 decades ago. In this review we provide an in-depth overview of the relevant topics involved in DNP-enhanced MAS NMR spectroscopy. This includes the theoretical description of DNP mechanisms as well as of the polarization transfer pathways that can lead to a uniform or selective spreading of polarization between nuclear spins. Furthermore, we cover historical and state-of-the art aspects of dedicated instrumentation, polarizing agents, and optimization techniques for efficient MAS DNP. Finally, we present an extensive overview on applications in the fields of structural biology and materials science, which underlines that MAS DNP has moved far beyond the proof-of-concept stage and has become an important tool for research in these fields.
The adaptive immune system is able to detect and destroy cells that are malignantly transformed or infected by intracellular pathogens. Specific immune responses against these cells are elicited by antigenic peptides that are presented on major histocompatibility complex class I (MHC I) molecules and recognized by cytotoxic T lymphocytes at the cell surface. Since these MHC I-presented peptides are generated in the cytosol by proteasomal protein degradation, they can be metaphorically described as a window providing immune cells with insights into the state of the cellular proteome. A crucial element of MHC I antigen presentation is the peptide-loading complex (PLC), a multisubunit machinery, which contains as key constituents the transporter associated with antigen processing (TAP) and the MHC I-specific chaperone tapasin (Tsn). While TAP recognizes and shuttles the cytosolic antigenic peptides into the endoplasmic reticulum (ER), Tsn samples peptides in the ER for their ability to form stable complexes with MHC I, a process called peptide proofreading or peptide editing. Through its selection of peptides that improve MHC I stability, Tsn contributes to the hierarchy of immunodominant peptide epitopes. Despite the fact that it concerns a key event in adaptive immunity, insights into the catalytic mechanism of peptide proofreading carried out by Tsn have only lately been gained via biochemical, biophysical, and structural studies. Furthermore, a Tsn homolog called TAP-binding protein-related (TAPBPR) has only recently been demonstrated to function as a second MHC I-specific chaperone and peptide proofreader. Although TAPBPR is PLC-independent and has a distinct allomorph specificity, it is likely to share a common catalytic mechanism with Tsn. This review focuses on the current knowledge of the multivalent protein–protein interactions and the concomitant dynamic molecular processes underlying peptide-proofreading catalysis. We do not only derive a model that highlights the common mechanistic principles shared by the MHC I editors Tsn and TAPBPR, and the MHC II editor HLA-DM, but also illustrate the distinct quality control strategies employed by these chaperones to sample epitopes. Unraveling the mechanistic underpinnings of catalyzed peptide proofreading will be crucial for a thorough understanding of many aspects of immune recognition, from infection control and tumor immunity to autoimmune diseases and transplant rejection.
In 1905, the managing editor of the Jewish Encyclopedia, Isidore Singer (1859–1939), published an article in the journal Ost und West from a "bird’s eye perspective on the development of American Jewry in the last 250 years." In this historical overview, Singer eventually attested that Jewish scholarship in America had an "absolute dependency on the European motherland." This judgment was based on his disapproving view of the two American rabbinical seminaries that existed at that time. According to Singer, there were still no scholars at the Hebrew Union College (HUC) in Cincinnati of the "already American[-born] generation of Israel." In fact, Singer’s observation was appropriate because it applied to the Jewish Theological Seminary of America (JTSA) in New York as much as to the HUC.3 Despite the history of Jewish settlement in America, around 1900 there was still no native Jewish scholarship in America. The scene was dominated by scholars educated in Europe, who often came with broken English and a strict academic sense of mission. In 1903, Kaufmann Kohler (1843–1926), born in Bavaria and trained at German universities, was chosen as the president of HUC. And a year earlier, Solomon Schechter (1847–1915) had been called to the JTSA in New York as its new president. ...
Proteins and glycolipids have been found to be decorated with phosphorylcholine (PC) both in protozoa and nematodes that parasitize humans and animals. PC epitopes can provoke various effects on immune cells leading to an immunomodulation of the host’s immune system that allows long-term persistence of the parasites. So far, only a limited number of PC-modified proteins, mainly from nematodes, have been identified. Infections caused by Leishmania spp. (e.g., L. infantum in southern Europe) affect about 12 million people worldwide and are characterized by a wide spectrum of clinical forms in humans, ranging from cutaneous to fatal visceral leishmaniasis. To establish and maintain the infection, these protozoa are dependent on the secretion of effector molecules into the host for modulating their immune system. In this project, we analyzed the PC modification of L. infantum promastigotes by 2D-gel based proteomics. Western blot analysis with the PC-specific antibody TEPC-15 revealed one PC-substituted protein in this organism, identified as eEF1α. We could demonstrate that the binding of eEF1α to one of its downstream effectors is dependent on its PC-modification. In this study we provide evidence that in this parasite the modification of eEF1α with PC may be essential for its function as an important virulence factor.
Motivated by a recent finding of an exact solution of the relativistic Boltzmann equation in a Friedmann–Robertson–Walker spacetime, we implement this metric into the newly developed transport approach Simulating Many Accelerated Strongly-interacting Hadrons (SMASH). We study the numerical solution of the transport equation and compare it to this exact solution for massless particles. We also compare a different initial condition, for which the transport equation can be independently solved numerically. Very nice agreement is observed in both cases. Having passed these checks for the SMASH code, we study a gas of massive particles within the same spacetime, where the particle decoupling is forced by the Hubble expansion. In this simple scenario we present an analysis of the freeze-out times, as function of the masses and cross sections of the particles. The results might be of interest for their potential application to relativistic heavy-ion collisions, for the characterization of the freeze-out process in terms of hadron properties.
Background: Although mortality after cardiac surgery has significantly decreased in the last decade, patients still experience clinically relevant postoperative complications. Among others, atrial fibrillation (AF) is a common consequence of cardiac surgery, which is associated with prolonged hospitalization and increased mortality.
Methods: We retrospectively analyzed data from patients who underwent coronary artery bypass grafting, valve surgery or a combination of both at the University Hospital Muenster between April 2014 and July 2015. We evaluated the incidence of new onset and intermittent/permanent AF (patients with pre- and postoperative AF). Furthermore, we investigated the impact of postoperative AF on clinical outcomes and evaluated potential risk factors.
Results: In total, 999 patients were included in the analysis. New onset AF occurred in 24.9% of the patients and the incidence of intermittent/permanent AF was 59.5%. Both types of postoperative AF were associated with prolonged ICU length of stay (median increase approx. 2 days) and duration of mechanical ventilation (median increase 1 h). Additionally, new onset AF patients had a higher rate of dialysis and hospital mortality and more positive fluid balance on the day of surgery and postoperative days 1 and 2. In a multiple logistic regression model, advanced age (odds ratio (OR) = 1.448 per decade increase, p < 0.0001), a combination of CABG and valve surgery (OR = 1.711, p = 0.047), higher C-reactive protein (OR = 1.06 per unit increase, p < 0.0001) and creatinine plasma concentration (OR = 1.287 per unit increase, p = 0.032) significantly predicted new onset AF. Higher Horowitz index values were associated with a reduced risk (OR = 0.996 per unit increase, p = 0.012). In a separate model, higher plasma creatinine concentration (OR = 2.125 per unit increase, p = 0.022) was a significant risk factor for intermittent/permanent AF whereas higher plasma phosphate concentration (OR = 0.522 per unit increase, p = 0.003) indicated reduced occurrence of this arrhythmia.
Conclusions: New onset and intermittent/permanent AF are associated with adverse clinical outcomes of elective cardiac surgery patients. Different risk factors implicated in postoperative AF suggest different mechanisms might be involved in its pathogenesis. Customized clinical management protocols seem to be warranted for a higher success rate of prevention and treatment of postoperative AF.
The etiology and pathogenesis of rheumatoid arthritis (RA) are marked by a complex interplay of various cell populations and is mediated by different signaling pathways. Traditionally, therapies have primarily focused on pain relief, reducing inflammation and the recovery of joint function. More recently, however, researchers have discussed the therapeutic efficacy of autologous platelet-rich plasma (PRP). The main objective of this work is to examine the influences of platelet-released growth factor (PRGF) on human synoviocytes under inflammatory conditions. Additionally, it is checked to which extend treatment with platelet concentrate influences the release of cytokines form synoviocytes. For this purpose, an in vitro RA model was created by stimulating the cells with the TNF-α. The release of cytokines was measured by ELISA. The cytokine gene expression was analyzed by real-time PCR. It has been observed that the stimulation concentration of 10 ng/ml TNF-α resulted in a significantly increased endogenous secretion and gene expression of IL-6 and TNF-α. The anti-inflammatory effect of PRGF could be confirmed through significant reduction of TNF-α and IL-1β. An induced inflammatory condition seems to cause PRGF to inhibit the release of proinflammatory cytokines. Further study is required to understand the exact effect mechanism of PRGF on synoviocytes.
The soluble loop BC region guides, but not dictates, the assembly of the transmembrane cytochrome b6

Studying folding and assembly of naturally occurring α-helical transmembrane proteins can inspire the design of membrane proteins with defined functions. Thus far, most studies have focused on the role of membrane-integrated protein regions. However, to fully understand folding pathways and stabilization of α–helical membrane proteins, it is vital to also include the role of soluble loops. We have analyzed the impact of interhelical loops on folding, assembly and stability of the heme-containing four-helix bundle transmembrane protein cytochrome b6 that is involved in charge transfer across biomembranes. Cytochrome b6 consists of two transmembrane helical hairpins that sandwich two heme molecules. Our analyses strongly suggest that the loop connecting the helical hairpins is not crucial for positioning the two protein “halves” for proper folding and assembly of the holo-protein. Furthermore, proteolytic removal of any of the remaining two loops, which connect the two transmembrane helices of a hairpin structure, appears to also not crucially effect folding and assembly. Overall, the transmembrane four-helix bundle appears to be mainly stabilized via interhelical interactions in the transmembrane regions, while the soluble loop regions guide assembly and stabilize the holo-protein. The results of this study might steer future strategies aiming at designing heme-binding four-helix bundle structures, involved in transmembrane charge transfer reactions.
Background: PINK1 deficiency causes the autosomal recessive PARK6 variant of Parkinson’s disease. PINK1 activates ubiquitin by phosphorylation and cooperates with the downstream ubiquitin ligase PARKIN, to exert quality control and control autophagic degradation of mitochondria and of misfolded proteins in all cell types.
Methods: Global transcriptome profiling of mouse brain and neuron cultures were assessed in protein-protein interaction diagrams and by pathway enrichment algorithms. Validation by quantitative reverse transcriptase polymerase chain reaction and immunoblots was performed, including human neuroblastoma cells and patient primary skin fibroblasts.
Results: In a first approach, we documented Pink1-deleted mice across the lifespan regarding brain mRNAs. The expression changes were always subtle, consistently affecting “intracellular membrane-bounded organelles”. Significant anomalies involved about 250 factors at age 6 weeks, 1300 at 6 months, and more than 3500 at age 18 months in the cerebellar tissue, including Srsf10, Ube3a, Mapk8, Creb3, and Nfkbia. Initially, mildly significant pathway enrichment for the spliceosome was apparent. Later, highly significant networks of ubiquitin-mediated proteolysis and endoplasmic reticulum protein processing occurred. Finally, an enrichment of neuroinflammation factors appeared, together with profiles of bacterial invasion and MAPK signaling changes—while mitophagy had minor significance. Immunohistochemistry showed pronounced cellular response of Iba1-positive microglia and GFAP-positive astrocytes; brain lipidomics observed increases of ceramides as neuroinflammatory signs at old age.
In a second approach, we assessed PINK1 deficiency in the presence of a stressor. Marked dysregulations of microbial defense factors Ifit3 and Rsad2 were consistently observed upon five analyses: (1) Pink1 −/− primary neurons in the first weeks after brain dissociation, (2) aged Pink1 −/− midbrain with transgenic A53T-alpha-synuclein overexpression, (3) human neuroblastoma cells with PINK1-knockdown and murine Pink1 −/− embryonal fibroblasts undergoing acute starvation, (4) triggering mitophagy in these cells with trifluoromethoxy carbonylcyanide phenylhydrazone (FCCP), and (5) subjecting them to pathogenic RNA-analogue poly(I:C). The stress regulation of MAVS, RSAD2, DDX58, IFIT3, IFIT1, and LRRK2 was PINK1 dependent. Dysregulation of some innate immunity genes was also found in skin fibroblast cells from PARK6 patients.
Conclusions: Thus, an individual biomarker with expression correlating to progression was not identified. Instead, more advanced disease stages involved additional pathways. Hence, our results identify PINK1 deficiency as an early modulator of innate immunity in neurons, which precedes late stages of neuroinflammation during alpha-synuclein spreading.

Es wird also die Binnenperspektive 'des Westens' dargelegt. Da das Anliegen dieses Artikels konzeptionell ist, wird darauf verzichtet, Quellenmaterial zu zitieren, stattdessen nur auf weiterführende Literatur verwiesen, die die Sachverhalte, auf die in der Analyse des Begriffs des Westens Bezug genommen wird, näher erläutern.
Disabling practices

Following Foucault’s theory of discourse this article aims at reformulating the established concept of disability. To this end, the author reconstructs ways in which disabling practices of subjectivation occur in and through public media discourses. The article focuses on the discoursive production of infantile identities in people with cognitive disabilities. The examples demonstrate that this discoursive production occurs in self-representational media formats and in outside media representations. Hence, the author develops a concept of disability as a discoursively produced ordering category, from which follows a reformulation of the disability concept. This reformulated concept, which grasps disability as discourse disability, allows in turn for a perspective on disability as practice and thus as independent from the subject. To conclude, the article discusses implications of such a perspective of disability for pedagogy and the social sciences, ultimately arguing for a broader definition of disability and for making respective benefits a matter of social pedagogy.
Establishing a yeast-based screening system for discovery of human GLUT5 inhibitors and activators

Human GLUT5 is a fructose-specific transporter in the glucose transporter family (GLUT, SLC2 gene family). Its substrate-specificity and tissue-specific expression make it a promising target for treatment of diabetes, metabolic syndrome and cancer, but few GLUT5 inhibitors are known. To identify and characterize potential GLUT5 ligands, we developed a whole-cell system based on a yeast strain deficient in fructose uptake, in which GLUT5 transport activity is associated with cell growth in fructose-based media or assayed by fructose uptake in whole cells. The former method is convenient for high-throughput screening of potential GLUT5 inhibitors and activators, while the latter enables detailed kinetic characterization of identified GLUT5 ligands. We show that functional expression of GLUT5 in yeast requires mutations at specific positions of the transporter sequence. The mutated proteins exhibit kinetic properties similar to the wild-type transporter and are inhibited by established GLUT5 inhibitors N-[4-(methylsulfonyl)-2-nitrophenyl]-1,3-benzodioxol-5-amine (MSNBA) and (−)-epicatechin-gallate (ECG). Thus, this system has the potential to greatly accelerate the discovery of compounds that modulate the fructose transport activity of GLUT5.
Background. Colorectal cancers (CRC) shed DNA into blood circulation. There is growing evidence that the analysis of circulating tumor DNA can be effectively used for monitoring of disease, to track tumor heterogeneity and to evaluate response to treatment. Case Presentation. Here, we describe two cases of patients with advanced CRC. The first case is about a patient with no available tissue for analysis of RAS mutation status. Liquid biopsy revealed RAS-wild-type and the therapy with anti-EGFR (epidermal growth factor receptor) monoclonal antibody cetuximab could be initiated. In the second case, the mutational profile of a patient with initial wild-type RAS-status was continually tracked during the course of treatment. An acquired KRAS exon 3 mutation was detected. The number of KRAS mutated fragments decreased continuously after the discontinuation of the therapy with EGFR-specific antibodies. Conclusion. Liquid biopsy provides a rapid genotype result, which accurately reproduces the current mutation status of tumor tissue. Furthermore, liquid biopsy enables close monitoring of the onset of secondary resistance to anti-EGFR therapy.
Working memory and conscious perception are thought to share similar brain mechanisms, yet recent reports of non-conscious working memory challenge this view. Combining visual masking with magnetoencephalography, we investigate the reality of non-conscious working memory and dissect its neural mechanisms. In a spatial delayed-response task, participants reported the location of a subjectively unseen target above chance-level after several seconds. Conscious perception and conscious working memory were characterized by similar signatures: a sustained desynchronization in the alpha/beta band over frontal cortex, and a decodable representation of target location in posterior sensors. During non-conscious working memory, such activity vanished. Our findings contradict models that identify working memory with sustained neural firing, but are compatible with recent proposals of ‘activity-silent’ working memory. We present a theoretical framework and simulations showing how slowly decaying synaptic changes allow cell assemblies to go dormant during the delay, yet be retrieved above chance-level after several seconds.
The adoption of a consistent alien species pathways categorization, hierarchy and terminology is crucial for increasing the interoperability of different online databases. In the present paper the European Alien Species Information Network (EASIN) classification system of pathways is compared and discussed with the classification scheme recently published by the Convention on Biological Diversity (CBD). Although the main pathway categories of the two classifications overall match, there are substantial differences in their subcategorization, with EASIN including 20 pathway subcategories while CBD considers 44 subcategories. In most cases, each EASIN subcategory pathway can correspond to two or more CBD subcategories. About 5,500 species listed in EASIN do not match directly with the CBD pathway subcategories, most of which are terrestrial invertebrates. Aiming at achieving synchronization between the two classification systems and at facilitating the access to information to researchers and policy makers, EASIN is trying to align with the CBD pathway classification scheme. This alignment process requires the involvement of a large number of experts, especially from the terrestrial realm, and the adoption of commonly accepted definitions of the CBD pathways.
Reactive oxygen species are produced transiently in response to cell stimuli, and function as second messengers that oxidize target proteins. Protein-tyrosine phosphatases are important reactive oxygen species targets, whose oxidation results in rapid, reversible, catalytic inactivation. Despite increasing evidence for the importance of protein-tyrosine phosphatase oxidation in signal transduction, the cell biological details of reactive oxygen species-catalyzed protein-tyrosine phosphatase inactivation have remained largely unclear, due to our inability to visualize protein-tyrosine phosphatase oxidation in cells. By combining proximity ligation assay with chemical labeling of cysteine residues in the sulfenic acid state, we visualize oxidized Src homology 2 domain-containing protein-tyrosine phosphatase 2 (SHP2). We find that platelet-derived growth factor evokes transient oxidation on or close to RAB5+/ early endosome antigen 1− endosomes. SHP2 oxidation requires NADPH oxidases (NOXs), and oxidized SHP2 co-localizes with platelet-derived growth factor receptor and NOX1/4. Our data demonstrate spatially and temporally limited protein oxidation within cells, and suggest that platelet-derived growth factor-dependent “redoxosomes,” contribute to proper signal transduction.
Background: Treatment of patients presenting with possible acute myocardial infarction (AMI) is based on timely diagnosis and proper risk stratification aided by biomarkers. We aimed at evaluating the predictive value of GDF-15 in patients presenting with symptoms suggestive of AMI.
Methods: Consecutive patients presenting with suspected AMI were enrolled in three study centers. Cardiovascular events were assessed during a follow-up period of 6 months with a combined endpoint of death or MI.
Results: From the 1818 enrolled patients (m/f = 1208/610), 413 (22.7%) had an acute MI and 63 patients reached the combined endpoint. Patients with MI and patients with adverse outcome had higher GDF-15 levels compared with non-MI patients (967.1pg/mL vs. 692.2 pg/L, p<0.001) and with event-free patients (1660 pg/mL vs. 756.6 pg/L, p<0.001). GDF-15 levels were lower in patients with SYNTAX score ≤ 22 (797.3 pg/mL vs. 947.2 pg/L, p = 0.036). Increased GDF-15 levels on admission were associated with a hazard ratio of 2.1 for death or MI (95%CI: 1.67–2.65, p<0.001) in a model adjusted for age and sex and of 1.57 (1.13–2.19, p = 0.008) adjusted for the GRACE score variables. GDF-15 showed a relevant reclassification with regards to the GRACE score with an overall net reclassification index (NRI) of 12.5% and an integrated discrimination improvement (IDI) of 14.56% (p = 0.006).
Conclusion: GDF-15 is an independent predictor of future cardiovascular events in patients presenting with suspected MI. GDF-15 levels correlate with the severity of CAD and can identify and risk-stratify patients who need coronary revascularization.
Despite the growth of Open Access, potentially illegally circumventing paywalls to access scholarly publications is becoming a more mainstream phenomenon. The web service Sci-Hub is amongst the biggest facilitators of this, offering free access to around 62 million publications. So far it is not well studied how and why its users are accessing publications through Sci-Hub. By utilizing the recently released corpus of Sci-Hub and comparing it to the data of ~28 million downloads done through the service, this study tries to address some of these questions. The comparative analysis shows that both the usage and complete corpus is largely made up of recently published articles, with users disproportionately favoring newer articles and 35% of downloaded articles being published after 2013. These results hint that embargo periods before publications become Open Access are frequently circumnavigated using Guerilla Open Access approaches like Sci-Hub. On a journal level, the downloads show a bias towards some scholarly disciplines, especially Chemistry, suggesting increased barriers to access for these. Comparing the use and corpus on a publisher level, it becomes clear that only 11% of publishers are highly requested in comparison to the baseline frequency, while 45% of all publishers are significantly less accessed than expected. Despite this, the oligopoly of publishers is even more remarkable on the level of content consumption, with 80% of all downloads being published through only 9 publishers. All of this suggests that Sci-Hub is used by different populations and for a number of different reasons, and that there is still a lack of access to the published scientific record. A further analysis of these openly available data resources will undoubtedly be valuable for the investigation of academic publishing.
Tumor-associated macrophages, angiogenesis, and tumor cell migration in oral squamous cell carcinoma

Objective: To investigate the relationship between tumor-associated macrophages (TAMs), neovascularization, and tumor cell migration in oral squamous cell carcinoma (OSCC) of an African subpopulation.
Materials and Methods: Twenty OSCC paraffin blocks underwent immunohistochemistry to TAM1 (CCR7), TAM2 (CD206), Twist, E-cadherin, N-cadherin, and CD34. The relative percentage of CCR7 + and CD206 + cells to overall immune cell population was calculated for three high power fields and an average was taken. TAM-related microvessel density (MVD) was determined as the mean of the three recorded values. Cases that had no CD34 + vessels adjacent to the TAMs region were regarded as having an MVD score of 0.
Results: Ten cases (50%) expressed greater CCR7 activity than CD206, seven cases (35%) expressed approximately equal activity of CCR7 and CD206, while three cases (15%) expressed greater activity of CD206 than CCR7. Twist expression was strong in some cases with strong N-cadherin and weak E-cadherin, but the expression of Twist was not consistently high in all cases that expressed strong N-cadherin and weak E-cadherin.
Conclusions: TAMs distribution suggested antitumor activity and the potential for tumor metastasis was only partly due to Twist-mediated epithelial–mesenchymal transition.
Drug product performance testing is an important part of quality-by-design approaches, but this process often lacks the underlying mechanistic understanding of the complex interactions between the disintegration and dissolution processes involved. Whereas a recent draft guideline by the US Food and Drug Administration (FDA) has allowed the replacement of dissolution testing with disintegration testing, the mentioned criteria are not globally accepted. This study provides scientific justification for using disintegration testing rather than dissolution testing as a quality control method for certain immediate release (IR) formulations. A mechanistic approach, which is beyond the current FDA criteria, is presented. Dissolution testing via United States Pharmacopeial Convention Apparatus II at various paddle speeds was performed for immediate and extended release formulations of metronidazole. Dissolution profile fitting via DDSolver and dissolution profile predictions via DDDPlus™ were performed. The results showed that Fickian diffusion and drug particle properties (DPP) were responsible for the dissolution of the IR tablets, and that formulation factors (eg, coning) impacted dissolution only at lower rotation speeds. Dissolution was completely formulation controlled if extended release tablets were tested and DPP were not important. To demonstrate that disintegration is the most important dosage form attribute when dissolution is DPP controlled, disintegration, intrinsic dissolution and dissolution testing were performed in conventional and disintegration impacting media (DIM). Tablet disintegration was affected by DIM and model fitting to the Korsmeyer–Peppas equation showed a growing effect of the formulation in DIM. DDDPlus was able to predict tablet dissolution and the intrinsic dissolution profiles in conventional media and DIM. The study showed that disintegration has to occur before DPP-dependent dissolution can happen. The study suggests that disintegration can be used as performance test of rapidly disintegrating tablets beyond the FDA criteria. The scientific criteria and justification is that dissolution has to be DPP dependent, originated from active pharmaceutical ingredient characteristics and formulations factors have to be negligible.
Obsessive-compulsive disorder (OCD) is a common neuropsychiatric disease affecting about 2% of the general population. It is characterized by persistent intrusive thoughts and repetitive ritualized behaviors. While gene variations, malfunction of cortico-striato-thalamo-cortical (CSTC) circuits, and dysregulated synaptic transmission have been implicated in the pathogenesis of OCD, the underlying mechanisms remain largely unknown. Here we show that OCD-like behavior in mice is caused by deficiency of SPRED2, a protein expressed in various brain regions and a potent inhibitor of Ras/ERK-MAPK signaling. Excessive self-grooming, reflecting OCD-like behavior in rodents, resulted in facial skin lesions in SPRED2 knockout (KO) mice. This was alleviated by treatment with the selective serotonin reuptake inhibitor fluoxetine. In addition to the previously suggested involvement of cortico-striatal circuits, electrophysiological measurements revealed altered transmission at thalamo-amygdala synapses and morphological differences in lateral amygdala neurons of SPRED2 KO mice. Changes in synaptic function were accompanied by dysregulated expression of various pre- and postsynaptic proteins in the amygdala. This was a result of altered gene transcription and triggered upstream by upregulated tropomyosin receptor kinase B (TrkB)/ERK-MAPK signaling in the amygdala of SPRED2 KO mice. Pathway overactivation was mediated by increased activity of TrkB, Ras, and ERK as a specific result of SPRED2 deficiency and not elicited by elevated brain-derived neurotrophic factor levels. Using the MEK inhibitor selumetinib, we suppressed TrkB/ERK-MAPK pathway activity in vivo and reduced OCD-like grooming in SPRED2 KO mice. Altogether, this study identifies SPRED2 as a promising new regulator, TrkB/ERK-MAPK signaling as a novel mediating mechanism, and thalamo-amygdala synapses as critical circuitry involved in the pathogenesis of OCD.
