This paper describes a method to treat contextual equivalence in polymorphically typed lambda-calculi, and also how to transfer equivalences from the untyped versions of lambda-calculi to their typed variant, where our specific calculus has letrec, recursive types and is nondeterministic. An addition of a type label to every subexpression is all that is needed, together with some natural constraints for the consistency of the type labels and well-scopedness of expressions. One result is that an elementary but typed notion of program transformation is obtained and that untyped contextual equivalences also hold in the typed calculus as long as the expressions are well-typed. In order to have a nice interaction between reduction and typing, some reduction rules have to be accompanied with a type modification by generalizing or instantiating types.
Background Multimorbidity is a highly frequent condition in older people, but well designed longitudinal studies on the impact of multimorbidity on patients and the health care system have been remarkably scarce in numbers until today. Little is known about the long term impact of multimorbidity on the patients' life expectancy, functional status and quality of life as well as health care utilization over time. As a consequence, there is little help for GPs in adjusting care for these patients, even though studies suggest that adhering to present clinical practice guidelines in the care of patients with multimorbidity may have adverse effects. Methods The study is designed as a multicentre prospective, observational cohort study of 3.050 patients aged 65 to 85 at baseline with at least three different diagnoses out of a list of 29 illnesses and syndromes. The patients will be recruited in approx. 120 to 150 GP surgeries in 8 study centres distributed across Germany. Information about the patients' morbidity will be collected mainly in GP interviews and from chart reviews. Functional status, resources/risk factors, health care utilization and additional morbidity data will be assessed in patient interviews, in which a multitude of well established standardized questionnaires and tests will be performed. Discussion The main aim of the cohort study is to monitor the course of the illness process and to analyse for which reasons medical conditions are stable, deteriorating or only temporarily present. First, clusters of combinations of diseases/disorders (multimorbidity patterns) with a comparable impact (e.g. on quality of life and/or functional status) will be identified. Then the development of these clusters over time will be analysed, especially with regard to prognostic variables and the somatic, psychological and social consequences as well as the utilization of health care resources. The results will allow the development of an instrument for prediction of the deterioration of the illness process and point at possibilities of prevention. The practical consequences of the study results for primary care will be analysed in expert focus groups in order to develop strategies for the inclusion of the aspects of multimorbidity in primary care guidelines.
Background: The standard electrode array for the MED-EL MAESTRO cochlear implant system is 31 mm in length which allows an insertion angle of approximately 720°. When fully inserted, this long electrode array is capable of stimulating the most apical region of the cochlea. No investigation has explored Electrically Evoked Compound Action Potential (ECAP) recordings in this region with a large number of subjects using a commercially available cochlear implant system. The aim of this study is to determine if certain properties of ECAP recordings vary, depending on the stimulation site in the cochlea. Methods: Recordings of auditory nerve responses were conducted in 67 subjects to demonstrate the feasibility of ECAP recordings using the Auditory Nerve Response Telemetry (ART™) feature of the MED-EL MAESTRO system software. These recordings were then analyzed based on the site of cochlear stimulation defined as basal, middle and apical to determine if the amplitude, threshold and slope of the amplitude growth function and the refractory time differs depending on the region of stimulation. Results: Findings show significant differences in the ECAP recordings depending on the stimulation site. Comparing the apical with the basal region, on average higher amplitudes, lower thresholds and steeper slopes of the amplitude growth function have been observed. The refractory time shows an overall dependence on cochlear region; however post-hoc tests showed no significant effect between individual regions. Conclusions :Obtaining ECAP recordings is also possible in the most apical region of the cochlea. However, differences can be observed depending on the region of the cochlea stimulated. Specifically, significant higher ECAP amplitude, lower thresholds and steeper amplitude growth function slopes have been observed in the apical region. These differences could be explained by the location of the stimulating electrode with respect to the neural tissue in the cochlea, a higher density, or an increased neural survival rate of neural tissue in the apex. Background Public health systems are confronted with constantly rising costs. Furthermore, diagnostic as well as treatment services become more and more specialized. These are the reasons for an interdisciplinary project on the one hand aiming at simplification of planning and scheduling patient appointments, on the other hand at fulfilling all requirements of efficiency and treatment quality. Methods As to understanding procedure and problem solving activities, the responsible project group strictly proceeded with four methodical steps: actual state analysis, analysis of causes, correcting measures, and examination of effectiveness. Various methods of quality management, as for instance opinion polls, data collections, and several procedures of problem identification as well as of solution proposals were applied. All activities were realized according to the requirements of the clinic's ISO 9001:2000 certified quality management system. The development of this project is described step by step from planning phase to inauguration into the daily routine of the clinic and subsequent control of effectiveness. Results Five significant problem fields could be identified. After an analysis of causes the major remedial measures were: installation of a patient telephone hotline, standardization of appointment arrangements for all patients, modification of the appointments book considering the reason for coming in planning defined working periods for certain symptoms and treatments, improvement of telephonic counselling, and transition to flexible time planning by daily updates of the appointments book. After implementation of these changes into the clinic's routine success could be demonstrated by significantly reduced waiting times and resulting increased patient satisfaction. Conclusion Systematic scrutiny of the existing organizational structures of the outpatients' department of our clinic by means of actual state analysis and analysis of causes revealed the necessity of improvement. According to rules of quality management correcting measures and subsequent examination of effectiveness were performed. These changes resulted in higher satisfaction of patients, referring colleagues and clinic staff the like. Additionally the clinic is able to cope with an increasing demand for appointments in outpatients' departments, and the clinic's human resources are employed more effectively.
In dieser Arbeit wurden zwei Systeme der biologischen Energiewandlung mit verschiedenen spektroskopischen Methoden untersucht und es wurden neue Erkenntnisse über die Funktion und Aktivierung der Proteine Proteorhodopsin und RuBisCO gewonnen. Zusätzlich konnte eine neue methodische Herangehensweise zur Untersuchung von Carboxylierungsreaktionen etabliert werden. Dieser Ansatz bietet in Zukunft breite Anwendungsmöglichkeiten zur Studie dieser biologisch so bedeutenden Reaktionsklasse. Mit Hilfe der Infrarotspektroskopie und vor allem durch den Einsatz von Tieftemperaturmessungen konnte der bisher kontrovers diskutierte Photozyklus von Proteorhodopsin (PR) eingehend charakterisiert werden. Jenseits des gut verstandenen aktiven Transports bei pH 9,0 wurde vor allem der pH 5,1 Photozyklus untersucht. Erstmals konnte auch in Infrarotspektren das M-Intermediat bei pH 5,1 nachgewiesen werden. Dieses Intermediat ist von entscheidender Bedeutung für den aktiven Transport über die Zellmembran und seine Existenz wurde bisher vielfach angezweifelt. Zudem konnte Glu-108 als ein möglicher Protonenakzeptor des Photozyklus bei pH 5,1 identifiziert werden. Durch einen pH-Indikator ließ sich der Nachweis erbringen, dass auch im sauren pH-Bereich Protonen freigesetzt werden. Damit steht fest, dass ein aktiver Protonentransport bei pH 5,1 möglich ist. Zusammen mit Informationen zu protonierbaren Aminosäureseitenketten (vornehmlich Asp und Glu) lässt sich zudem mit Einschränkungen die These unterstützen, dass PR ober- und unterhalb des pKa-Werts von Asp-97 in verschiedene Richtungen Protonen pumpt. Damit ergibt sich ein differenziertes Bild für den pH-abhängigen Photozyklus von PR mit drei pH-Bereichen (pH 9,0, 8,5 bis 5,5 und 5,1) in denen PR unterschiedliche Protonentransportwege zeigt. Als weiteres biologischen System wurde RuBisCO genauer untersucht. Im Fokus der Arbeit war dabei die Aktivierung durch die Bildung eines Lysin-Carbamats im aktiven Zentrum. Obwohl RuBisCO das am häufigsten vorkommende Enzym unseres Planeten ist, in der Kohlenstofffixierung eine bedeutende Rolle spielt und obwohl mehrere Dutzend Kristallstrukturen existieren, gibt es noch immer genügend offene Fragen zur Aktivierung. Mit Hilfe eines Käfig-CO2 konnte die Carbamatbildung im Enzym direkt verfolgt und der Einfluss von Magnesiumionen auf die Aktivierung beobachtet werden. Damit ließ sich ganz klar ausschließen, dass Magnesium bereits für die Carbamatbildung erforderlich ist. Die Koordination von Mg2+ ist erst für die Endiol-Bildung im weiteren Reaktionszyklus essentiell. Zusätzlich wurde gezeigt, dass Azid eine Inhibierung des Enzyms durch die Konkurrenz mit CO2 um die Bindungsstelle auslöst, allerdings verdrängt CO2 das Azidion im Laufe der Zeit. Mit den Ergebnissen für RuBisCO konnte klar gezeigt werden, dass die Kombination aus Käfig-CO2 und Rapid-Scan IR-Spektroskopie ein völlig neues Feld für die Untersuchung von Carboxylierungsreaktionen eröffnet. Gerade die offenen Fragen zu Biotin bindenden Carboxylasen bieten ein breites Anwendungsgebiet für diese Methodik.
This study addresses the structure-function relationships of three essential membrane proteins: Porin from Paracoccus denitrificans, Porin OmpG from Eschericia coli and BetP from Corynobacterium glutamicum using Fourier transform infrared (FT-IR) spectroscopy and Attenuated Total Reflection (ATR) techniques. The structure of porin from P. denitrificans is known for more than a decade; however, the mechanism for loss of functionality together with the monomerization was not clear. In this study we have addressed the role of lipids for the functionality of porin using FT-IR. OmpF porin was found to interact with the lipid molecules via the aromatic girdles surrounding the protein for functionality. In this study, molecular bonds and groups of the lipids were established as reporter groups probing at different depths of the bilayer in order to understand the interaction partner of the aromatic girdles of porins. Monomerization of the trimeric assembly of OmpF porin reconstituted in lipids is induced by increasing the temperature. Porin (OmpF) was found to be extremely stable: The secondary structure of the protein was unaltered up to the temperature-induced main transition, around 80-90 °C, above which it is denatured. However, the interaction of the aromatic girdle with the lipid molecules exhibited distinct changes at much lower temperature values (40 - 50°) where, according to the previous functional studies, monomerization and the loss of function occurs. The results are compared with OmpG porin from E.coli, for which the functional unit is a monomer. The aromatic girdle-lipid interaction was monitored by the tyrosine aromatic ring C=C vibrational mode, a universal marker for the protein stability and interaction. We have also found that the aromatic girdles of porins are interacting with the interfacial region of the lipid bilayer instead of lipid headgroups. Lipid-protein interaction was found to be not only essential for the structural stability, but also for the functionality of OmpF porin. We have also studied the structural properties of OmpG from E.coli. The structure of OmpG at two pH values has been resolved using X-ray crystallography and the channel has been proposed to attain different states at different pH values as closed (pH < 5.5) and open (pH >7.5). This study, using IR spectroscopy, revealed that the pH-induced opening and closing of the channel is reflected by the frequency shifts of the ? sheet structure. OmpG has more rigid ? barrel properties upon opening of the channel. IR spectral analysis revealed multiple ? sheet signals with different hydrogen bond strengths. This enabled us to monitor the formation of hydrogen bridges between the extracellular loops upon opening of the channel. The conclusion that OmpG porin having two states at different pH values was also confirmed by the three mutants where the role of the histidine pair (H231 & H261) and loop 6 has been addressed. Temperature-profiling of the wild type (WT) protein and the mutants did not show pH dependent structural stability differences in detergent solution. However, the WT protein was found to be more stable in the open form in 2D crystals than the closed form. Reconstitution into lipids has increased the transition temperature value by ~20 °C in the closed state and ~25 °C in the open state. Therefore we conclude that the open and closed state of OmpG has structural stability differences that are only revealed in the lipid environment. A comparison of the transition temperature values of OmpG WT and the mutants suggested that the hydrogen bond network among S218-H231-H261-D267, together with the formation of 12 residue-long ?-sheet contributes to the structural stability of the open channel. In the process of closing and opening of the channel, the globular structure of the protein remains mainly unchanged, while there are changes in the side chain moieties. In addition to the role of the histidine pair and the loop L6, in situ opening/closing experiments showed that the negatively charged amino acids, i.e. Asp and Glu, and Arg residues also play an active role; possibly by interacting with each other inside the pore lumen. Therefore it could be concluded that the closure of the channel at acidic pH values is not only via closing the channel entrance by loop 6, but also via changing the electric potential inside the lumen due to the different states of charged amino acids in order to effectively block the gateway. BetP from C.glutamicum attains an active and inactive state in order to adjust its glycine betaine uptake rate to the osmotic conditions that the cell encounters. The structure of BetP is not yet available. The WT protein exhibited structural differences in the presence of excess K+, which is one of the activation conditions. In 2D crystals, increasing the ionic strength to 700 mM K+ was shown to induce changes in the ?-helical moiety with contributions from the ester groups and one Tyr residue using ATR-FTIR. An increase in ionic strength to 220 mM K+ was found to be the threshold value of potassium concentration ([K+]) where the protein exhibits structural alterations in detergent solution. The determined [K+] values are in good agreement with the previous functional studies. However, there are differences in the activation profile of BetP in 2D crystals and in detergent solution, which points out that the lipids are involved in the conformational transition from the inactive to the active state and their absence can lead to different structural properties. BetP WT was found to have ~65% alpha-helix, ~25% random coil and ~10% turn structure in detergent solution. In the presence of excess K+, the WT protein is found to adapt more unordered structure. Secondary structure analysis of the mutants revealed that both the N- and C-terminus are in ?-helical conformation. Reconstitution of WT protein in 2D crystals increased the main transition (denaturation) temperature value from ~62 °C to ~85 °C, a clear indication that the protein is more stable in lipid environment. Temperature-profiling of the two forms of the WT protein revealed that the structural breakdown is preceeded by monomerization of the trimeric assembly. Comparing the two forms of the WT protein and the mutant BetA, we conclude that the oligomeric status is stabilized via the interactions among hydrophilic regions involving the N terminus. H/D exchange and activation with excess K+ in D2O-buffer revealed that activation of the protein involves the interaction of Arg and Asp/Glu residues in the cytoplasmic region of the protein. BetP WT and the two mutants tested, i.e. BetA and BetP?C45, showed differences in protein packing upon activation. The WT protein and BetP?C45 mutant also show changes in the hydrogen bonding properties of turns. Since BetA does not show such a property in activation, we conclude that the N-terminus interacts with the loops in the inactive state via the interaction of charged amino acids for the WT protein and that this interaction is altered during the activation. It could be argued that the protein packing is affected via the changes in turns upon activation. We also have found experimental evidence that one Tyr residue has different orientations in the active and inactive state of BetP. Based on the previous functional studies, it could be one of the five Tyr residues in the cytoplasmic region of the protein (in loop 3, 6, 7 or C-terminus). The mutant BetP?C45, on the other hand, showed fewer differences between the active and inactive state conditions and based on the H/D exchange rates, the mutant shows the properties of an active WT protein, proving that the C-terminal truncation impairs the conformational transition between the active and inactive states.
Die in den 1990er Jahren erneuerte sozialwissenschaftliche Debatte um Staatsbürgerschaft (citizenship) als ein Verhältnis von Nation, Staat und Individuum, das über rein rechtliche Arrangements hinausgeht und auch Fragen der politischen und kulturellen Zugehörigkeit im weiteren Sinn betrifft, hat seit fünf bis zehn Jahren auch die Literatur- und Kulturwissenschaften erreicht, mit eigenen Schwerpunktsetzungen und Konzepten. Eines dieser Konzepte, das des sexual citizenship, ist das Thema der kulturwissenschaftlichen Magisterarbeit "We Will Be Citizens: The Notion of Citizenship in Tony Kushner's Angels in America and Larry Kramer's The Normal Heart". Kramer's The Normal Heart (1985) und Kuschner's Angels in America (1993/95) werden als Beispiele für Theaterstücke der 'ersten' und 'zweiten' Generation, die sich mit der AIDS-Krise der 1980er Jahre auseinandersetzen analysiert; dabei, so das zentrale Argument, geht es in beiden Stücken darum, die Auseinandersetzung mit AIDS und die formulierte Kritik an der unzureichenden Reaktion der Reagan-Administration auf die AIDS-Krise im Kontext von Neuverhandlungen von sexual citizenship zu lesen, also als Neuverhandlung der Rechte und gesellschaftlichen Teilhabemöglichkeit für als von der 'sexuellen Norm abweichend' definierte Personengruppen, im Kontext der Stücke vor allem homosexuelle Männer. Dabei wird citizenship klar als ein kulturelles Konzept definiert und macht dadurch auch die Bedeutung kultureller Produktionen (z.B. Literatur) für die Debatten deutlich: "Citizenship is not only to be understood as a political issue, but also as socially and materially constructed and culturally coded. If we understand culture as the ways of doing things and of organizing society, then the negotiations of citizenship take place, among other ways, through culture" (5). Die Diskussion des Themas beginnt mit einer zielgerichteten Skizze der theoretischen Debatten um sexual citizenship, der Rolle von AIDS für diese Debatten (Kap. 2) und einer Übersicht über die AIDS-Krise der 1980er Jahre (Kap. 3). Dabei stellt er mit Sontag, Yingling und Isin die Konstruktionen von Alterität – krank/gesund, self/other, citizen/non-citizen – die die Diskussion prägen, in den Vordergrund der Analyse. In den beiden folgenden Kapitel (4 und 5) werden die beiden Stücke mit Blick auf die jeweilige Aushandlung von sexual citizenship diskutiert; für Kramers Stück wird dabei das Thema schwule Identität, eine auf Aktivismus hin ausgerichtete Agenda und das Plädoyer für gesellschaftliche Anerkennung hervorgestellt, während bei der Analyse von Kuschners Text dessen Inszenierung von 'Geschichte' und ideologischer Kritik im Vordergrund steht. Den Abschluss bildet ein Vergleich der beiden Stücke in Hinsicht auf ihre jeweilige Agenda und deren Umsetzung. The Normal Heart, verfolgt insgesamt eine eher assimilatorische Sto߬richtung, die versucht, Homosexualität zu 'norm(alis)ieren', während Angels, radikaler, die Konstruktionen von sexuellen Normen hinterfragt und ein Recht auf Differenz proklamiert (z.B. S. 69).
Biophysikalische Charakterisierung von endogenen Ionenkanälen (P2X7, TRPV) in humanen Mastzellen

Die Akupunktur ist zentraler Bestandteil der traditionellen ostasiatischen Medizin, die auch Moxibustion und Kräuterheilkunde (Herbalmedizin) umfasst [Focks et al. 2006]. Akupunkturpunkte sind auch durch eine höhere Konzentration von sensorischen Rezeptoren und Mastzellen charakterisiert [Dung 1984; Heine 1988; Hwang 1992]. So ergaben Untersuchungen, dass die Stimulation (physikalische und chemische Reize) von Akupunkturpunkten auf Rezeptoren und auch Mastzellen einwirkt [Belmonte 1996; Schmidt 2002; Yang et al. 2007; Zhang et al. 2008a; Zhang et al. 2008b; Zhao 2008]. Dabei zeigten auch Pflanzenkomponenten aus der TCM Einflüsse [Lee 2000; Smith et al. 2006]. ...
Bacterial autotransporters represent a diverse family of proteins that autonomously translocate across the inner membrane of Gram-negative bacteria via the Sec complex and across the outer bacterial membrane. They often possess exceptionally long N-terminal signal sequences. We analyzed 90 long signal sequences of bacterial autotransporters and members of the two-partner secretion pathway in silico and describe common domain organization found in 79 of these sequences. The domains are in agreement with previously published experimental data. Our algorithmic approach allows for the systematic identification of functionally different domains in long signal sequences. Methanosarcina acetivorans kann Kohlenmonoxid (CO) als Kohlenstoff- und Energiequelle nutzen. Als Endprodukte entstehen bei der Verwertung von CO neben Methan signifikante Mengen an Azetat und Formiat sowie Dimethylsulfid (DMS). In dieser Arbeit sollten verschiedene Aspekte dieses außergewöhnlichen CO-Stoffwechsels analysiert werden. Folgende Ergebnisse wurden erzielt: 1) Weder die Methanogenese, noch die Bildung eines der anderen Metaboliten wird durch hohe CO-Partialdrücke gehemmt. Inhibitorstudien mit BES belegen, dass die CO-Oxidation und die Bildung von Azetat, Formiat und DMS nicht an die Methanogenese gekoppelt sind. Inhibitorstudien legen nahe, dass die Methanogenese aus CO am Aufbau eines Na+-Gradienten beteiligt ist und das Vorhandensein eines vom Protonenpotential-abhängigen Schrittes. 2) Eine neue, kostengünstige Transformationsmethode mittels Polyethylenglykol (PEG) konnte für M. acetivorans etabliert werden. Die Transformationshäufigkeit betrug ca. 1,1 x 107 Transformanden/μg DNA und liegt damit im Bereich von der der bisher etablierten Liposomen-vermittelten Transformationsmethode. 3) Mutantenanalysen und physiologische Studien belegen eine Beteiligung der Mts-Proteine in der DMS-Bildung und DMS-Verwertung, da in ihrer Abwesenheit kein DMS aus CO gebildet, kein Methan aus DMS produziert wird, und M. acetivorans nicht mehr auf DMS als Energiequelle wachsen kann. Die Mts-Proteine sind für das carboxidotrophe Wachstum jedoch nicht essentiell. Immunologische Analysen belegen eine substratabhängige Regulation von MtsF und weisen auf genetische Interaktionen der einzelnen Loci oder der Isoformen selbst hin. 4) Die monofunktionellen CODH-Isoformen von M. acetivorans sind am carboxidotrophen Wachstum beteiligt, jedoch nicht essentiell. Die beiden Isoformen der bifunktionellen CODH/ACS sind funktionell, und wenigstens eine von ihnen ist für autotrophes als auch carboxidotrophes Wachstum notwendig. Eine mögliche posttranslationale Modifikation von Cdh1 weist auf unterschiedliche physiologische Funktion und/oder Lokalisation hin. 5) Die F420H2-Dehydrogenase ist essentiell für methylotrophes, nicht jedoch für carboxidotrophes Wachstum.
Deichbau und andere flussbautechnische Maßnahmen haben dazu geführt, dass die Mittlere Elbe ihre ursprünglichen Überschwemmungsgebiete verloren hat. Um die Auswirkungen der alljährlich auftretenden Hochwasserereignisse einzudämmen, wurden große Bereiche der Talniederung durch Deiche vom Überflutungsgeschehen abgetrennt. Diese Eingriffe in den Naturhaushalt ermöglichten gleichfalls eine intensive ackerbauliche Nutzung oder eine hochwassersichere Bebauung der Auen. Die natürliche Auendynamik ist heute weitestgehend auf einen schmalen Bereich entlang der Elbe beschränkt. Hinter den Deichen sind die für die Elbeauen typischen Lebensräume von der lebenswichtigen Auendynamik abgeschnitten. Angepasste Auenarten und -lebensgemeinschaften treten zugunsten von Allerweltsarten zurück. Eine Wiederanbindung von Altauenbereichen an das Überflutungsgeschehen ist deshalb eine der vordringlichsten Maßnahmen zur Revitalisierung gefährdeter Auenlebensräume und stellt eine Chance dar, einen nachhaltigen und modernen Hochwasserschutz mit Naturschutzzielen zu verbinden. An der Elbe entspricht das aktuelle Hochwasserschutzsystem nicht den heutigen Anforderungen an den Hochwasserschutz. Um jedoch jederzeit auf mögliche große Hochwasserereignisse reagieren zu können, entstanden Anfang der 1990er Jahre in den Anliegerländern der Elbe zahlreiche Pläne für Deichrückverlegungen.
Motivated by the question of correctness of a specific implementation of concurrent buffers in the lambda calculus with futures underlying Alice ML, we prove that concurrent buffers and handled futures can correctly encode each other. Correctness means that our encodings preserve and reflect the observations of may- and must-convergence, and as a consequence also yields soundness of the encodings with respect to a contextually defined notion of program equivalence. While these translations encode blocking into queuing and waiting, we also describe an adequate encoding of buffers in a calculus without handles, which is more low-level and uses busy-waiting instead of blocking. Furthermore we demonstrate that our correctness concept applies to the whole compilation process from high-level to low-level concurrent languages, by translating the calculus with buffers, handled futures and data constructors into a small core language without those constructs.
In seinem 2005 mit dem Booker-Preis ausgezeichneten Roman The Sea nimmt sich der irische Schriftsteller John Banville dem Thema Trauma, Erinnerung und Trauer durch den Einsatz intermedialer Manifestationen, gestalterisch an. Das Werk schildert vor allem die subtilen Auswirkungen eines Kindheitstraumas auf die Identitätsbildung und Lebensführung des alternden Protagonisten, dessen Wahrnehmungsweise und Weltanschauung durch das Medium der bildenden Kunst geprägt sind.
Tongruben sind bedeutende Sekundärstandorte seltener und gefährdeter Pflanzenarten sowie bemerkenswerter Pflanzengesellschaften. Charakteristische, darunter gefährdete Arten, die in Tongruben Nordrhein-Westfalens nachgewiesen werden konnten, werden genannt. Aussagen über den naturschutzfachlichen Wert und die Entwicklung derartiger Biotope werden diskutiert. Rekultivierungsarbeiten sollten sich deshalb auf Flächen beschränken, die Bodenbefestigung erfordern. Zudem sollte höchstens bodenständiges Saat- und Pflanzgut verwendet werden.
»Wenn es einen Wirklichkeitssinn gibt, dann muß es« – so folgerte Robert Musil zu Beginn des 20. Jahrhunderts – »auch einen Möglichkeitssinn geben.« Darunter versteht er die Fähigkeit, »alles, was ebenso gut [auch] sein könnte, zu denken und das, was ist, nicht wichtiger zu nehmen, als das, was nicht ist.« Mit dem Begriff des Möglichkeitssinns, der auf die Relativität und Alternativität des individuellen Denkens sowie auf die Utopie eines anderen, hypothetischen Lebens verweist, hat Robert Musil in seinem Jahrhundertroman Der Mann ohne Eigenschaften dem Kontingenzbewusstsein des modernen Menschen Ausdruck gegeben, welches am Ende des 20. Jahrhunderts zum Grundmodus der Existenz und der Verfasstheit des Individuums überhaupt werden sollte. Dem Begriff der Kontingenz liegt bei aller Unschärfe ein grundlegendes, auf Aristoteles zurückgehendes Verständnis zugrunde, welches Niklas Luhmann folgendermaßen definiert: Kontingent ist etwas, was weder notwendig ist, noch unmöglich ist; was also so, wie es ist (war, sein wird), sein kann, aber auch anders möglich ist. Der Begriff bezeichnet mithin Gegebenes (zu Erfahrendes, Erwartetes, Gedachtes, Phantasiertes) im Hinblick auf mögliches Anderssein; er bezeichnet Gegenstände im Horizont möglicher Abwandlungen.
Durch die auffällig aufgewölbten, mehr oder weniger halbkugeligen, im feuchten Zustand etwas glänzend hell bläulich-grünen Polster, die sich bei Trockenheit weiß färben (deutscher und wissenschaftlicher [Gattungs-]Name!) und im unteren Teil abgestorben gelblich-bräunlich sind, ist dieses Moos eine der bekanntesten Laubmoosarten, auch wenn seine Einzelstämmchen für sich betrachtet gar nicht so spektakulär wirken. Aber sie sind lang: Im Durchschnitt erreichen sie 10 cm, aber können unter günstigen Bedingungen problemlos doppelt so lang werden - und entsprechend hoch sind dann die Polster. Diese liegen oft isoliert oder in kleinen Gruppen auf dem Waldboden - wie abgefallen oder weggeworfen zwischen Laub und geraten schnell wegen ihrer Form und Farbe ins Blickfeld.
In previous investigations an impact of cellular copper homeostasis on ageing of the ascomycete Podospora anserina has been demonstrated. Here we provide new data indicating that mitochondria play a major role in this process. Determination of copper in the cytosolic fraction using total reflection X-ray fluorescence spectroscopy analysis and eGfp reporter gene studies indicate an age-related increase of cytosolic copper levels. We show that components of the mitochondrial matrix (i.e. eGFP targeted to mitochondria) become released from the organelle during ageing. Decreasing the accessibility of mitochondrial copper in P. anserina via targeting a copper metallothionein to the mitochondrial matrix was found to result in a switch from a copper-dependent cytochrome-c oxidase to a copper-independent alternative oxidase type of respiration and results in lifespan extension. In addition, we demonstrate that increased copper concentrations in the culture medium lead to the appearance of senescence biomarkers in human diploid fibroblasts (HDFs). Significantly, expression of copper-regulated genes is induced during in vitro ageing in medium devoid of excess copper suggesting that cytosolic copper levels also increase during senescence of HDFs. These data suggest that the identified molecular pathway of age-dependent copper dynamics may not be restricted to P. anserina but may be conserved from lower eukaryotes to humans.
The title compound, C25H22O5, was obtained by a dehydrogenative carbonylation reaction. It crystallizes with one half-molecule in the asymmetric unit. The molecules have crystallographic C2 symmetry and the two atoms of the carbonyl group are located on the rotation axis. The methoxy groups are coplanar with the benzene ring to which they are attached [C-C-O-C = 1.0 (6)°]. The two furan rings are inclined at 17.3 (3)° with respect to each other and the dihedral angle between the furan ring and the benzene ring is 75.83 (12)°. The crystal structure is stabilized by C-H...O hydrogen bonds. The Mg centre in the title compound, [MgBr2(C2H7N)3], is pentacoordinated in a trigonal-bipyramidal mode with the two Br atoms in axial positions and the N atoms of the dimethylamine ligands in equatorial positions. The MgII centre is located on a crystallographic twofold rotation axis. The crystal structure is stabilized by N—H⋯Br hydrogen bonds. The N atom and H atoms of one dimethylamine ligand are disordered over two equally occupied positions.
Der Aufenthalt in einem Krankenhaus beginnt in der Regel mit der präklinischen Versorgung des Patienten durch den Rettungsdienst. Hier ist die Schnittstelle zwischen nichtärztlicher und ärztlicher Versorgung. Umso wichtiger ist hier ein reibungsloser und vor allem komplikationsloser Verlauf. Aus diesem Grund wurde, basierend auf Ergebnissen dieser Studie, eine Schulung zur fehlerfreien und effizienten Kommunikation für Rettungsassistenten entwickelt. Sie beinhaltete Hintergrundwissen, fünf klare Kommunikationsregeln (Schaffen einer eindeutigen Sprache, Setzen von Prioritäten, Bilden von Kommunikationsschleifen, Abschaffen eines „übertriebenen Wir-Gefühls“, Bestimmen eines Teamführers) und das BAUM-Schema zur Patientenübergabe. BAUM ist ein Akronym für Bestand, Anamnese, klinische Untersuchungsergebnisse und getroffene Maßnahmen. Fragestellung: Lassen sich durch eine kurze Schulung und die Einführung des BAUM-Schemas die Kommunikation in einem Rettungsdienstteam und die Patientenübergabe zwischen Rettungsdienstmitarbeitern und Notarzt verbessern? Methoden: In der vorliegenden Studie bildeten simulierte Notfallszenarien, die von zwei Rettungsassistenten zu bearbeiten waren, die Basis. Es wurde die Kommunikation zwischen den Teammitgliedern und das jeweilige Patientenübergabegespräch vor (Projektstufe 1) und nach einer Schulung (Projektstufe 2) evaluiert. Dabei gab es vier verschiedene Arten von Notfallszenarien; drei wurden von einem echten Patientendarsteller gespielt (Asthma bronchiale, Lungenembolie, Polytrauma) und eines mit einer Simulationspuppe dargestellt (kardiopulmonale Reanimation). Jedes Rettungsdienstteam hatte einen Fall in einem realitätsnahen Umfeld zu bearbeiten. Die Szenarien wurden digital aufgezeichnet und mit den dafür entwickelten Analysewerkzeugen evaluiert. Dabei wurde auf negative Kommunikationsereignisse, wie beispielsweise „Missverständnisse“ oder „Kompetenzgerangel“, geschaut und der Informationsfluss von Rettungsassistent zum Notarzt beobachtet. Die Schulung nach der ersten Projektstufe erfolgte in drei Unterrichtseinheiten und wurde interaktiv gestaltet; die aktive Mitarbeit durch das Bearbeiten von Arbeitsblättern sollte möglichst viele Lerntypen ansprechen. Im Anschluss an jede Schulung wurden Fragen diskutiert und Übungsaufgaben bearbeitet. Alle Probanden waren Rettungsassistenten der Berufsfeuerwehr Frankfurt am Main und nahmen nur ein einziges Mal an einer Simulation teil. Die Patientenschauspieler mussten ausreichende medizinische Kenntnisse vorweisen. Ergebnisse: In der ersten Projektstufe wurden 20 Szenarien simuliert, in der zweiten 14. Somit wurden insgesamt 68 Rettungsassistenten und 34 Übergaben evaluiert. Auf ein ausgewogenes Verhältnis der einzelnen Notfallszenarien wurde geachtet. Die Gesamtzahl der aufgezeichneten negativen Kommunikationsereignisse pro simuliertem Szenario wurde von 3,9 auf 1,4 vermindert (p=0,003). Gleichzeitig wurden die einzelnen negativen Kommunikationsereignisse pro Notfall ausgewertet. Durch die Schulung sollten sie in Projektstufe 2 vermindert werden. Hier ergab sich aber kein eindeutiger Trend zur Veränderung. Dagegen steigerte sich nach Einführung des BAUM-Schemas der Informationsgehalt in allen Übergaben von Rettungsassistenten zum Notarzt von 52,6 % auf 67,8 % (p=0,021), beim Szenario Lungenembolie wurde sogar eine Steigerung von 77,1 % erreicht. Konklusion: Die kurzen Schulungen zur Kommunikation während eines Rettungsdiensteinsatzes blieben in ihrer Effizienz hinter den Erwartungen zurück. Hier muss davon ausgegangen werden, dass andere Faktoren wie unterschiedliche Vorkenntnisse der Probanden bezüglich der Kommunikationsfähigkeit die Ursache waren. Die Patientenübergabe nach Einführung des BAUM-Schemas konnte dagegen nachweislich verbessert werden. Es führte nicht nur zu einer inhaltlichen Vervollständigung, sondern bot dem Rettungsassistenten gleichzeitig eine Gliederung zur Wiedergabe. Somit war eine sinnvolle Strukturierung der Informationen über den Patienten erreichbar. Es wurde damit dem Notarzt ermöglicht, mehr Informationen über den Patienten zu erhalten und mehr über die Lage am Notfallort zu erfahren.
This thesis is devoted to the developement of a classical model for the study of the energetics and stability of carbon nanotubes. The motivation behind such a model stems from the fact that production of nanotubes in a well-controlled manner requires a detailed understanding of their energetics. In order to study this different theoretical approaches are possible, ranging from the computationally expensive quantum mechanical first principle methods to the relatively simple classical models. A wisely developed classical model has the advantage that it could be used for systems of any possible size while still producing reasonable results. The model developed in this thesis is based on the well-known liquid drop model without the volume term and hence we call it liquid surface model. Based on the assumption that the energy of a nanotube can be expressed in terms of its geometrical parameters like surface area, curvature and shape of the edge, liquid surface model is able to predict the binding energy of nanotubes of any chirality once the total energy and the chiral indices of it are known. The model is suggested for open end and capped nanotubes and it is shown that the energy of capped nanotubes is determined by five physical parameters, while for the open end nanotubes three parameters are sufficient. The parameters of the liquid surface model are determined from the calculations performed with the use of empirical Tersoff and Brenner potentials and the accuracy of the model is analysed. It is shown that the liquid surface model can predict the binding energy per atom for capped nanotubes with relative error below 0.3% from that calculated using Brenner potential, corresponding to the absolute energy difference being less than 0.01 eV. The influence of the catalytic nanoparticle on top of which a nanotube grows, on the nanotube energetics is also discussed. It is demonstrated that the presence of catalytic nanoparticle changes the binding energy per atom in such a way that if the interaction of a nanotube with the catalytic nanoparticle is weak then attachment of an additional atom to a nanotube is an energetically favourable process, while if the catalytic nanoparticle nanotube interaction is strong , it becomes energetically more favourable for the nanotube to collapse. The suggested model gives important insights in the energetics and stability of nanotubes of different chiralities and is an important step towards the understanding of nanotube growth process. Young modulus and curvature constant are calculated for single-wall carbon nanotubes from the paremeters of the liquid surface model and demonstrated that the obtained values are in agreement with the values reported earlier both theoretically and experimentally. The calculated Young modulus and the curvature constant were used to conclude about the accuracy of the Tersoff and Brenner potentials. Since the parameters of the liquid surface model are obtained from the Tersoff and Brenner potential calculations, the agreement of elastic properties derived from these parameters corresponds to the fact that both potentials are capable of describing the elastic properties of nanotubes. Finally, the thesis discuss the possible extension of the model to various systems of interest.
Hintergrund: Die Notwendigkeit der Etablierung der Anwendung von Evidenzbasierter Medizin (EbM) in den klinischen Alltag sowie die Sicherung der europaweiten Qualifikation in EbM ist seit geraumer Zeit vielfältig diskutiert worden. Gefördert durch das "Leonardo da Vinci"- Berufsbildungsprogramm der Europäischen Union hat das euebm-Projekt zum Ziel, durch ein europaweit einheitliches integriertes Basiscurriculum für evidenzbasierte Medizin eine bessere Verzahnung zwischen dem Erlernen der EbM-Techniken und der eigenen Patientenversorgung zu erreichen. An der Universitätsklinik Frankfurt am Main wird ab dem 01.01.2009 das euebm-Projekt in den klinischen Alltag integriert werden. Die Zielgruppe dieses Projektes bilden alle Assistenzärzte jeglicher Fachrichtungen im 1. Weiterbildungsjahr. Durch einen online-basierten Kurs wird den Interessenten die Möglichkeit geboten, an den einzelnen Lehrmodulen teilzunehmen.
Methoden: Es werden alle Assistenzärzte im 1. Weiterbildungsjahr in einem individualisierten Schreiben zur Teilnahme an dem Projekt aufgefordert. Nach der elektronischen Anmeldung werden die Teilnehmer prätest-Evaluationsbogen mit inhaltlichen EbM-Fragen beantworten. Danach durchlaufen die Teilnehmer ein EbM-Lernprogramm, bestehend aus 5 Modulen. Jedes Modul wird durch eine Lernerfolgskontrolle abgeschlossen. Abschließend wird eine Gesamtlernkontrolle von jedem Teilnehmer abverlangt. Am Ende des online-Kurses werden die Teilnehmer ebenfalls einen mehrteiligen subjektiven Evaluationsbogen beantworten. Um einen längerfristigen Lerneffekt messen zu können, werden 6 Monate bzw. 1 Jahr nach dem erfolgreichen Abschluss die Teilnehmer erneut einen objektiven inhaltlichen Evaluationsbogen bearbeiten.
Ergebnisse: Im Rahmen der 10. Jahrestagung des DNEbM im März 2009 können voraussichtlich Zwischenergebnisse der objektiven (präinterventionell vs. postinterventionell) und subjektiven Evaluationen veröffentlicht werden. Zudem wird derzeit die Erstellung einer statistischen Auswertung der Zugriffszahlen auf die verschiedenen medizinischen Online-Datenbanken und Suchmaschinen (prä- und postinterventionell) aufgebaut.
Schlussfolgerung/Implikation: Mit Hilfe der curricular vermittelten Lehrinhalte und Techniken sollen Ärzte in die Lage versetzt werden, EbM praxisnah für und mit ihren Patienten umzusetzen. In der konkreten Anwendung von EbM an ihren individuellen Patienten können Ärzte möglicherweise erfahren, dass die Integration von EbM in den klinischen Alltag ihr Spektrum an Beratungs- und Behandlungsmöglichkeiten erweitert.
Elektronentransferprozesse in Pd-Komplexen 1,4-Chinon-basierter Bis(pyrazol-1-yl)methan-Liganden

PdII-katalysierte Oxidationsreaktionen organischer Verbindungen stellen wichtige synthetische Werkzeuge dar. Um das Übergangsmetall in katalytischen Mengen einsetzen zu können ist es nötig, das im Zuge der Substratumwandlung entstehende Pd0 seinerseits wieder zu reoxidieren. Im großtechnischen Wacker-Prozess setzt man zu diesem Zweck das Redoxsystem CuII/CuI ein, welches Elektronen von Pd0 aufnimmt und sie auf molekularen Sauerstoff überträgt, so dass als einziges Abfallprodukt Wasser entsteht. Da beim Einsatz von Kupfersalzen allerdings Probleme durch unerwünschte Nebenreaktionen auftreten können, stieß man auf der Suche nach alternativen Redox-Cokatalysatoren auf 1,4-Benzochinon. Man nimmt an, dass der Reoxidation von Pd0 eine Koordination des 1,4-Benzochinons an das Übergangsmetallatom vorausgeht. Vor diesem Hintergrund wurde in der vorliegenden Arbeit ein 1,4-Naphthochinon-basierter Bis(pyrazol-1-yl)methan-Ligand entwickelt, dessen Pd-Komplex das katalytisch aktive Übergangsmetallatom und den Redox-Cokatalysator in einem einzigen Molekül vereint. Eine direkte intra- und intermolekulare 1,4-Chinon/Pd-Koordination wird durch das Ligandendesign jedoch verhindert. Anhand dieses Modellsystems sollte dann untersucht werden, ob alternativ zu den etablierten mechanistischen Vorstellungen zum Ablauf des Elektronentransfers, eine durch den Raum verlaufende elektronische Kommunikation der beiden redoxaktiven Zentren möglich ist. Sollte das der Fall sein, wäre man künftig bei der Entwicklung und Optimierung solcher Hybridkatalysatoren nicht auf die Miteinbeziehung des 1,4-Chinons in die Koordinationssphäre des Übergangsmetallions angewiesen, sondern könnte diese ausschließlich an die koordinationschemischen Erfordernisse des Pd-Zentrums anpassen. Nachdem das Modellsystem zur Verfügung stand, galt es im ersten Schritt, die elektrochemischen Eigenschaften von freiem Ligand und PdII-Komplex mittels Cyclovoltammetrie zu bestimmen. Mit Kenntnis der Redoxpotentiale wurden daraufhin elektrochemisch selektiv reduzierte Spezies des Liganden bzw. seines PdII-Komplexes erzeugt und deren UV-vis-Spektren in situ aufgenommen. Letztere lieferten nötige Referenzwerte für die folgenden Experimente. Auf Grundlage dieser Vorarbeiten konnte nun die elektronische Kommunikation der redoxaktiven Zentren in dem Komplex untersucht werden. Da im Modellsystem zunächst beide redoxaktive Zentren in der oxidierten Form vorliegen, war es erforderlich, das PdII-Ion selektiv zu reduzieren, um dem System an der richtigen Stelle Elektronen zuzuführen. Als Reduktionsmittel wurde Triethylamin gewählt; entsprechende Vergleichsexperimente stellten zuvor sicher, dass dieses keine Nebenreaktionen mit dem Liganden selbst eingeht. Im Zuge der Reduktion des PdII-Zentrums ist eine Änderung der UV-vis-spektroskopischen Eigenschaften des Liganden zu beobachten. Ein Vergleich der erhaltenen Daten mit den Referenzspektren der elektrochemisch erzeugten reduzierten Spezies legt den Schluss nahe, dass nach Überführung des PdII-Zentrums in die nullwertige Stufe der durch den Raum verlaufende Übergang eines Elektrons auf den 1,4-Naphthochinon-basierten Liganden erfolgt. Das hierbei entstehende 1,4-Naphthosemichinonradikal wurde zusätzlich ESR-spektroskopisch nachgewiesen.
In the title compound, C11H14O4, an intermediate for the synthesis of a new kind of estrogen receptor modulator, all non-H atoms lie on a common plane (r.m.s. deviation = 0.0472 Å). All C-C bonds in the side chain are in a trans conformation, and the hydroxyl group is also trans to the methylene chain. In the crystal structure, molecules form centrosymmetric dimers showing a head-to-head arrangement which is stabilized by O-H...O hydrogen bonds. A weak C-H...O contact is also present.
Höhere Eukaryoten stellen ein Ensemble von Zellen dar, die in Kompartimente unterteilt sind. Somit sind intra- und interzelluläre Transportprozesse entscheidend für das Überleben dieser Zellverbände. In meiner Arbeit habe ich Evolution und Struktur von Translokationskomplexen untersucht, um einige Aspekte dieser komplexen Systeme zu untersuchen. Eingangs befassten wir uns mit Rezeptorsystemen am Beispiel des Proteintransports. Mittels phylogenetischer Analysen fanden wir heraus, dass Pex5 nicht der Urahn der anderen untersuchten 3-TPR-Domänen ist, obwohl Pex5 in allen eukaryotischen Organismen vorkommt. Ein Vergleich der 3-TPR-Domänen mit der restlichen Sequenz des Rezeptorproteins ergab, dass die 3-TPR-Domänen eine langsamere Evolutionsgeschwindigkeit aufweisen, was für eine Evolutionseinschränkung durch Interaktionspartner spricht. Sec72 ist möglicherweise aus einer TPR1 (Hop) Domäne entstanden und eine Funktion als Hsp70-erkennende Komponente des Sec-Komplexes für den post-translationalen Import kann daraus abgeleitet werden. „Recycling“ von 3-TPR-Domänen anderer Proteine konnten wir durch unsere phylogenetische Analyse auch für die zweite 3-TPR-Domäne von Tom34 nachweisen, die mit CYP40/FKBP51/52 clustert. Darüber hinaus war es uns möglich, die plastidär bzw. mitochondriell lokalisierten Formen von Toc64 phylogenetisch zu unterscheiden. Durch Erzeugung von Homologiemodellen konnten organellspezifische Aminosäuren strukturell eingeordnet werden. Dabei stellten wir fest, dass sich fast alle Positionen, die sich in der Aminosäurekomposition unterscheiden, auf der konvexen Seite der 3-TPR-Domäne befinden. Molekulardynamische Simulationen zeigten zudem deutliche Veränderung der Hauptbewegungen der 3-TPR-Domänen nach Komplexierung mit dem Hsp90-C-Terminus. Bei Bindung des Liganden werden intramolekulare Wasserstoffbrücken sowohl auf der konvexen als auch konkaven Seite der 3-TPR-Domäne „umgeschaltet“. Diese Erkenntnisse führen zu zwei Hypothesen: 1.) die Organellspezifität der Rezeptoren wird durch die Interaktion mit anderen Komplexpartnern garantiert und 2.) die Änderungen des Wasserstoffbrückennetzwerkes auf der konvexen Seite nach Hsp90-Bindung führen zur Ausbildung der Bindungsstelle für die andere Komplexkomponente. Beide Hypothesen erklären die experimentellen Beobachtungen bezüglich der Rezeptoren und warum keine phylogenetischen Hinweise für die Existenz von Vorstufenprotein-spezifischen Hsp70/90-Proteinen gefunden werden konnten. Nach dem Rezeptor haben wir uns mit dem Translokationsprozess befasst. Wir konnten phylogenetisch zeigen, dass sich Omp85 aus Proteobakterien im Vergleich zu Cyanobakterien und Eukaryoten insbesondere durch andersartige POTRA Domänen auszeichnet und fanden zwei konservierte Motive in der Porenregion. Zudem konnten wir im Heterokontophyten P. tricornutum ein vollständiges Omp85 identifizieren (bipartite Signalsequenz, 2 POTRAs, Pore mit langen Schleifen). Die Aminosäuresequenz weicht teils deutlich von den bekannten Omp85-Proteinen ab, was die Entdeckung erschwerte. Wir haben damit geklärt, dass auch im Translokationsapparat von komplexen Plastiden ein b-Fassprotein der Omp85 Familie die Kerneinheit bildet. Ebenfalls zu den Protein-transportierenden b-Fassproteinen gehört TolC, das aber im Gegensatz zu Omp85 auch andere Substanzen, wie zum Beispiel Siderophore transportiert. Alr2887 ist das einzige TolC-ähnliche Protein aus Anabaena sp. PCC7120. Vergleichende Phänotypuntersuchungen weisen auf eine Interaktion eines ABC-Transporters (DevBCA Operon) mit Alr2887 hin. Die Distanz zwischen äußerer Membran und Plasmamembran ist in Anabaena doppelt so groß wie in E. coli. Entsprechend fanden wir im Adapterprotein DevB eine stark verlängerte dimere Doppelwendel, die das von TolC gebildete a-Fass im Periplasma bis hin zum ABC-Transporter in der Plasmamembran theoretisch fortsetzen kann. Da verschiedenste in Anabaena existierende ABC-Transporter TolC als Abflusskanal benötigen, nehmen wir an, dass Alr2887 ein Rundumtalent in Bezug auf die zu transportierenden Substrate darstellt. Dieses ist auch aufgrund der basalen Einordnung im phylogenetischen Baum zu vermuten; es könnte somit auch in den „Multi-Drug-Efflux“ involviert sein. Nicht nur ABC-Transporter, auch TonB-abhängige Transporter stehen in funktionellem Zusammenhang mit TolC. Wir haben Aminosäuresequenzen von ~4600 TBDTs aus Gram-negativen Bakterien und Cyanobakterien zusammengetragen und nach ihrer paarweisen Ähnlichkeit geclustert. Anhand experimentell charakterisierter TBDTs mit bekannten Substraten und TBDTs mit vorhergesagten Substraten konnten wir sehr vielen Clustern ein Substrat zuordnen, das die in ihnen zusammengefassten TBDTs aller Wahrscheinlichkeit nach importieren. Wir konnten ferner feststellen, dass es noch eine Menge weiterer Cluster mit unbekannten Substratspezifitäten gibt und unsere Analysen stimulieren somit die Arbeiten an diesem System im Allgemeinen und in Cyanobakterien im Besonderen.
Background Different iron transport systems evolved in Gram-negative bacteria during evolution. Most of the transport systems depend on outer membrane localized TonB-dependent transporters (TBDTs), a periplasma-facing TonB protein and a plasma membrane localized machinery (ExbBD). So far, iron chelators (siderophores), oligosaccharides and polypeptides have been identified as substrates of TBDTs. For iron transport, three uptake systems are defined: the lactoferrin/transferrin binding proteins, the porphyrin-dependent transporters and the siderophore-dependent transporters. However, for cyanobacteria almost nothing is known about possible TonB-dependent uptake systems for iron or other substrates. Results We have screened all publicly available eubacterial genomes for sequences representing (putative) TBDTs. Based on sequence similarity, we identified 195 clusters, where elements of one cluster may possibly recognize similar substrates. For Anabaena sp. PCC 7120 we identified 22 genes as putative TBDTs covering almost all known TBDT subclasses. This is a high number of TBDTs compared to other cyanobacteria. The expression of the 22 putative TBDTs individually depends on the presence of iron, copper or nitrogen. Conclusions We exemplified on TBDTs the power of CLANS-based classification, which demonstrates its importance for future application in systems biology. In addition, the tentative substrate assignment based on characterized proteins will stimulate the research of TBDTs in different species. For cyanobacteria, the atypical dependence of TBDT gene expression on different nutrition points to a yet unknown regulatory mechanism. In addition, we were able to clarify a hypothesis of the absence of TonB in cyanobacteria by the identification of according sequences.
Um eine kardiale Ursache für den Zustand klinisch instabiler Patienten auszuschließen, gilt der Einsatz einer fokussierten Echokardiographie als aussagekräftige orientierende Untersuchungsmaßnahme. Auch während einer kardiopulmonalen Reanimation kann eine ALS-konforme Echokardiographie eingesetzt werden. Dafür benötigt der Notfallmediziner ein standardisiertes Training, welches spezielle Schwerpunkte beinhaltet. Dabei muss neben der Vermittlung von theoretischen Kenntnissen und der praktischen Anwendung der Echokardiographie auch besonders auf das Trainieren von visuell perzeptiven Fähigkeiten geachtet werden, da ein schnelles Erkennen von spezifischen Diagnosen elementar für die Prognose klinisch instabiler Patienten ist. Diese Arbeit befasst sich mit der Entwicklung und Anwendung eines computerbasierten Testverfahrens, der dem Erfassen des Lernerfolges im Rahmen des Stufe-1-Training für Notfallechokardiographie. Dabei wird vor allem in Hinblick auf die schnelle Identifikation von Blickdiagnosen geachtet, indem eine Zeitmessung erfolgte. Bei der Entwicklung wurden, basierend auf die aktuellen Empfehlungen für den Einsatz von Echokardiographie in der Intensiv-und Notfallmedizin, die 10 wichtigsten Echokardiographiediagnosen als Filmsequenzen dargestellt. Dazu gehören Befunde wie die PEA, der Perikarderguss, Asystolie, reduzierte Pumpfunktion und Rechtsherzbelastung. Zu jeder Filmsequenz, die je nach Version 5 Sekunden oder bis zu 60 Sekunden dauern, wurde eine Freitexteingabe gefordert und eine Multiple-Choice-Aufgabe gestellt. Ergebnisse wurden im Rahmen von drei Kurskonzepten für Stufe-1-Noallechokardiographie gesammelt. Damit einhergehend wurde die Validität des Simulationstests Notfallechokardiographie bewiesen. Bei der Betrachtung der Ergebnisse zeigt sich, dass es möglich ist, mit dem Test den Wissensstand der Teilnehmer aufzuzeigen. Dabei wurde deutlich, dass Befunde wie die PEA, der Perikarderguss und die hochgradig eingeschränkte Pumpfunktion von der Mehrzahl der Teilnehmer korrekt in Freitexteingabe und Multiple-Choice-Aufgabe benannt werden konnten. Schwächen im Erkennen bestimmter Befunde, allen voran des Normalbefundes, wurden anhand der Testanwendung aufgedeckt. Somit kann der Simulationstest Notfallechokardiographie zu einer Überarbeitung der Kursinhalte beitragen. Bei einer Erfassung der Zeit, die für die Identifikation der Befunde benötigt wird, wird ersichtlich, dass definierte Diagnosen innerhalb von 10 Sekunden erkannt werden können. Vor allem der Befund der PEA wurde nach dem Kurs schnell erkannt. Andere Diagnosen, wie das normale Herz, zeigen, dass es nach einem Kursprogramm weiterhin schwierig sein kann, bestimmte Befunde innerhalb von 10 Sekunden zu stellen. Die gewonnenen Ergebnisse werden durch die erhobenen Daten bei der Anwendung des Simulationstest Notfallechokardiographie in der 5-Sekunden-Version unterstrichen. Spannend ist, dass im Rahmen eines spezialisierten Trainings kein Unterschied in den Ergebnissen festzustellen war. Es spielte dabei keine Rolle, ob die Teilnehmer die Filmsequenzen 5 Sekunden oder 60 Sekunden sehen konnten. Aus diesen Ergebnissen lässt sich der Schluss ziehen, dass nach einem achtstündigen Kurs für ALS-konforme Echokardiographie definierte Blickdiagnosen dass definierte Befunde innerhalb 5 Sekunden erkannt werden können.
Die Thorax-, Trachea- und Lungensonographie in der Intensiv- und Notfallmedizin hat wegen der bettseitigen Anwendbarkeit einen hohen Stellenwert. Als „hand-held“ Verfahren eignet sich der Ultraschall für die Beurteilung von Pathologien während zeitkritischer Szenarien in Notaufnahmen, auf der Intensivstation sowie in der Präklinik. Auch Interventionen wie Pleurapunktion oder die perkutane Dilatationstracheotomie können sonographiegesteuert sicherer durchgeführt werden. Die wichtigste Voraussetzung hierfür ist jedoch, dass der Untersucher in der Anwendung der Sonographie ausgebildet wurde. Ein einheitliches, funktionierendes und wissenschaftlich validiertes Kurskurrikulum lag für diesen Bereich bisher aber nicht vor. Deshalb ist das Kurskonzept zur Thorax-, Trachea- und Lungensonographie in der Intensiv- und Notfallmedizin THOLUUSE entwickelt und die Frage gestellt worden, ob Ultraschallnovizen durch ein praxisorientiertes Lernsystem die Sonographie von Thorax, Trachea und Lunge innerhalb eines Lerntages adäquat erlernen und anwenden können. Das Kurrikulum richtet sich nach den geforderten Inhalten und den Stufenkonzepten der WHO, DEGUM und EFSUMB. Es berücksichtigt Anatomie, sonographische Anatomie und Physiologie. Standarduntersuchungen und Anlotungspunkte der Lungensonographie. Ebenso ist die Darstellung der Trachea gelehrt worden. Erkrankungen von Notfall- und Intensivpatienten, die ultraschallgestützte Punktionen und die sonographischen Algorithmen zur Untersuchung von Thorax, Trachea und Lunge sind ebenfalls besprochen und an Modellen geübt worden. Der Kurs gliedert sich in theoretische und praktische Lerneinheiten. Die Teilnehmer haben theoretischen Unterricht und zwei praktische Hands-on-Trainingseinheiten erhalten. Hier konnten innerhalb eines zyklischen Lernsystems mit durchschnittlich 6 Stationen zu je 10 min. die besprochenen Themen praktisch geübt und erlernt werden. Das Verhältnis von Instruktor zu Teilnehmer hat bei 1:2 gelegen. Um den Kurs evaluieren zu können ist der Lernerfolg von 54 Kursteilnehmern, die an insgesamt 4 Kursen teilgenommen haben, vor und nach dem Kurs in Theorie und Simulation überprüft worden. Nach den Kursen hat ein praktischer Posttest stattgefunden, der ebenfalls evaluiert worden ist. Hierbei hat sich eine signifikante Verbesserung der Teilnehmer in allen geprüften Abschnitten ergeben. In der Theorie haben sich die Teilnehmer von 58% richtige Antworten auf 84% verbessern können und in der Simulation haben die Teilnehmer nach dem Kurs 80% richtige Antworten erreicht. Im praktischen Posttest konnten 95% richtige Anlotungen erreicht werden. Das bedeutet, dass THOLUUSE erfolgreich durchgeführt werden konnte und die Teilnehmer die Thorax-, Trachea- und Lungensonographie erlernen und sich im Gegensatz zu den Ausgangswerten signifikant verbessern konnten. Es konnte gezeigt werden, dass die Sonographie von Thorax, Trachea und Lunge innerhalb eines eintägigen Kursprogrammes zu erlernen ist.
Es war das Ziel dieser Untersuchung, die Güte der Behandlung im Stammzelltransplantationszentrum Frankfurt a.M. zu eruieren. Besonderer Wert wurde auf die Analyse von Infektionen und TRM gelegt. Dabei sollten zwei Zeiträume miteinander verglichen werden. Es wurde gefunden, dass trotz Zunahme des Risikoprofils der Patienten sowie der Schwere der Erkrankungen im Zeitraum II die Inzidenz der Infektionen sowie die Rate an TRM stabil blieb. Die bei der Auswertung der Daten aufgetretenen Schwierigkeiten hinsichtlich der Erfassung der Daten und hinsichtlich der Vielfalt der erhobenen Daten zeigte uns, dass es eventuell sinnvoll ist, eine Studie mit ähnlichen Endpunkten anhand verschiedener, definierter Patientenkollektive zu erheben. Leider ist es nicht möglich gewesen, unser Patientenkollektiv zum Beispiel nach Erkrankungen zu separieren, weil die Fallzahlen für die Erkrankungen zu gering gewesen wären. Desweiteren war ja das Ziel der Studie, alle Patienten einzubinden und ein repräsentatives Bild der Transplantationen in Frankfurt am Main zu schauen. Wir haben in unserer Studie den Riskscore angewandt, um die Effizienz herauszufinden. Wir konnten zeigen, dass der Riskscore durch ein objektiviertes Verfahren eine realistische Einschätzung der Komplikationen und Überlebenswahrscheinlichkeiten bietet und in Zukunft für alle zu transplantierenden Patienten zur Risikostratifizierung eingesetzt werden sollte. Anhand des geschätzen Risikos lässt sich zum Beispiel das Verfahren der Konditionierung (intensivere Chemotherapie), des Monitorings oder der zusätzlichen Behandlungsmöglichkeiten (Prophylaxe,DLI, Immunmodulatoren) ändern. Außerdem erleichtert der Riskscore die Vergleichbarkeit mit anderen Studien.
Nach einer erfolgten Stammzelltransplantion im Rahmen einer Leukämie sollte in regelmäßigen Abständen der hämatopoetische Chimärismus untersucht werden, da ansteigende autologe Anteile einem Rezidiv häufig voran gehen. [33, 35-37] Es wurde in den letzten Jahren beschrieben [50, 52, 54, 55, 81], dass Sequenzpolymorphismen (SPs) als hochempfindliche Marker für die Chimärismusanalyse fungieren können. Durch sie würde eine deutlich höhere Sensitivität erzielt werden, als mit der bisher verwendeten Methode, die Short Tandem Repeats als Marker zur Diskriminierung von Spender und Empfänger benutzt. Ziel dieser Arbeit war es, die Proben von Kindern, die nach einer ALL eine Stammzelltransplantation erhalten hatten, und die bereits mit der STR-Methode untersucht worden waren, mit der RT-PCR-Methode in Hinblick auf die in der Einleitung gestellten Fragen, erneut zu analysieren. Es ist in 96 % der Empfänger-/Spenderpaare möglich unter den 29 ausgewähltenten SPs mindestens einen geeigneten Marker zu finden und sicherlich wäre es möglich noch weitere Sequenz-Polymorphismen hinzu zu nehmen, falls die Informativität der 29 verwendeten nicht ausreichend ist. Es konnte in fast allen Experimenten eine Sensitivität von 0,1 % erreicht werden, mit zunehmender experimenteller Erfahrung immer zuverlässiger, so dass man inzwischen ein Experiment, bei dem diese Sensitivität nicht erreicht wird, wiederholen würde. Durch eine Vereinfachung der Methode mit einem optimierten Primerscreening, universellen Standardreihen und dem Einsatz von einer definierten Menge DNA-Lösung, die zumindest über einen weiten Bereich unabhängig ist von der enthaltenen Konzentration, lässt sich eine Laborroutine entwickeln, die ähnlich zeitaufwändig ist, wie der jetzige Goldstandard, die STR-Methode. Allerdings ist die RT-PCR-Methode derzeit noch deutlich teurer. In dieser Arbeit zeigt sich, dass die Real-Time PCR mit Sequenz Polymorphismen als genetische Marker eine sehr sensitive Methode zur Erfassung autologer Anteile darstellt und in der praktischen Anwendbarkeit mit der bisherigen PCR-Methode mit Short Tandem Repeats zur Differenzierung vergleichbar ist. Häufig lassen sich autologe Signale früher detektieren. Dadurch werden auch mehr Patienten als gefährdet eingestuft. Vor allem Patienten, die zweimal oder öfter in Folge einen gemischten Chimärismus von größer als 0,5 % aufweisen (und sich nicht in der Phase eines abnehmenden Chimärismus befinden) müssen genau beobachtet und engmaschig kontrolliert werden. Bei einer ansteigenden Dynamik ist es häufig sinnvoll, eine Immuntherapie einzuleiten. Nur bei zwei unserer Patienten verschwanden die autologen Signale von alleine wieder. Bei 50 % der Rezidivpatienten und bei 2 der 3 Patienten, die abgestoßen haben, sieht man mit der RT-PCR-Methode früher autologe Signale. Es wäre jetzt in einem nächsten Schritt nötig, bei einem ausreichend großen Patientenkollektiv beide Methoden parallel in einer prospektiven Studie miteinander zu vergleichen.

L1-CAM (L1 cell-adhesion molecule), or more simply L1, plays an important role in the progression of human carcinoma. Overexpression promotes tumour-cell invasion and motility, growth in nude mice and tumour metastasis. It is feasible that L1-dependent signalling contributes to these effects. However, little is known about its mechanism in tumour cells. We reported previously that L1 is cleaved by ADAM (a disintegrin and metalloprotease) and that the cytoplasmic part is essential for L1 function. Here we analysed more closely the role of proteolytic cleavage in L1-mediated nuclear signalling. Using OVMz carcinoma cells and L1-transfected cells as a model, we found that ADAM10-mediated cleavage of L1 proceeds in lipid raft and non-raft domains. The cleavage product, L1-32, is further processed by PS (presenilin)/gamma-secretase to release L1-ICD, an L1 intracellular domain of 28 kDa. Overexpression of dominantnegative PS1 or use of a specific gamma-secretase inhibitor leads to an accumulation of L1-32. Fluorescence and biochemical analysis revealed a nuclear localization for L1-ICD. Moreover, inhibition of ADAM10 and/or gamma-secretase blocks nuclear translocation of L1-ICD and L1-dependent gene regulation. Overexpression of recombinant L1-ICD mediates gene regulation in a similar manner to full-length L1. Our results establish for the first time that regulated proteolytic processing by ADAM10 and PS/gamma-secretase is essential for the nuclear signalling of L1 in human carcinoma cell lines. Background Since June 2002, revised regulations in Germany have required "Emergency Medical Care" as an interdisciplinary subject, and state that emergency treatment should be of increasing importance within the curriculum. A survey of the current status of undergraduate medical education in emergency medical care establishes the basis for further committee work. Methods Using a standardized questionnaire, all medical faculties in Germany were asked to answer questions concerning the structure of their curriculum, representation of disciplines, instructors' qualifications, teaching and assessment methods, as well as evaluation procedures. Results Data from 35 of the 38 medical schools in Germany were analysed. In 32 of 35 medical faculties, the local Department of Anaesthesiology is responsible for the teaching of emergency medical care; in two faculties, emergency medicine is taught mainly by the Department of Surgery and in another by Internal Medicine. Lectures, seminars and practical training units are scheduled in varying composition at 97% of the locations. Simulation technology is integrated at 60% (n=21); problem-based learning at 29% (n=10), e-learning at 3% (n=1), and internship in ambulance service is mandatory at 11% (n=4). In terms of assessment methods, multiple-choice exams (15 to 70 questions) are favoured (89%, n=31), partially supplemented by open questions (31%, n=11). Some faculties also perform single practical tests (43%, n=15), objective structured clinical examination (OSCE; 29%, n=10) or oral examinations (17%, n=6). Conclusion Emergency Medical Care in undergraduate medical education in Germany has a practical orientation, but is very inconsistently structured. The innovative options of simulation technology or state-of-the-art assessment methods are not consistently utilized. Therefore, an exchange of experiences and concepts between faculties and disciplines should be promoted to guarantee a standard level of education in emergency medical care.
Various concurrency primitives had been added to functional programming languages in different ways. In Haskell such a primitive is a MVar, joins are described in JoCaml and AliceML uses futures to provide a concurrent behaviour. Despite these concurrency libraries seem to behave well, their equivalence between each other has not been proven yet. An expressive formal system is needed. In their paper "On proving the equivalence of concurrency primitives", Jan Schwinghammer, David Sabel, Joachim Niehren, and Manfred Schmidt-Schauß define a universal calculus for concurrency primitives known as the typed lambda calculus with futures. There, equivalence of processes had been proved. An encoding of simple one-place buffers had been worked out. This bachelor’s thesis is about encoding more complex concurrency abstractions in the lambda calculus with futures and proving correctness of its operational semantics. Given the new abstractions, we will discuss program equivalence between them. Finally, we present a library written in Haskell that exposes futures and our concurrency abstractions as a proof of concept.
Novel host records and feeding behaviors are reported for five species in three genera of two cassidine beetle tribes, Hemisphaerotini and Imatidiini, from Ecuador. Carludovica Ruiz and Pav. (Cyclanthaceae) is reported as a new plant family and genus host for two species of Spaethiella Barber and Bridwell. Calathea G. Mey (Marantaceae) is reported as the first host record for Aslamidium capense (Herbst) and Calathea lutea Schult. and Calathea majestica (Linden) H. Kenn. are reported as the first host records for Aslamidium semicirculare (Olivier). Immature stages of Demotispa elaeicola (Aslam) are reported for the first time; larvae are external folivores and both larvae and pupae are solitary and lack exuvio-fecal shields. The adults and larvae of D. elaeicola feed by rasping palm fruits, a rare feeding pattern in Cassidinae.
Background The to date evidence for a dose-response relationship between physical workload and the development of lumbar disc diseases is limited. We therefore investigated the possible etiologic relevance of cumulative occupational lumbar load to lumbar disc diseases in a multi-center case-control study. Methods In four study regions in Germany (Frankfurt/Main, Freiburg, Halle/Saale, Regensburg), patients seeking medical care for pain associated with clinically and radiologically verified lumbar disc herniation (286 males, 278 females) or symptomatic lumbar disc narrowing (145 males, 206 females) were prospectively recruited. Population control subjects (453 males and 448 females) were drawn from the regional population registers. Cases and control subjects were between 25 and 70 years of age. In a structured personal interview, a complete occupational history was elicited to identify subjects with certain minimum workloads. On the basis of job task-specific supplementary surveys performed by technical experts, the situational lumbar load represented by the compressive force at the lumbosacral disc was determined via biomechanical model calculations for any working situation with object handling and load-intensive postures during the total working life. For this analysis, all manual handling of objects of about 5 kilograms or more and postures with trunk inclination of 20 degrees or more are included in the calculation of cumulative lumbar load. Confounder selection was based on biologic plausibility and on the change-in-estimate criterion. Odds ratios (OR) and 95% confidence intervals (CI) were calculated separately for men and women using unconditional logistic regression analysis, adjusted for age, region, and unemployment as major life event (in males) or psychosocial strain at work (in females), respectively. To further elucidate the contribution of past physical workload to the development of lumbar disc diseases, we performed lag-time analyses. Results We found a positive dose-response relationship between cumulative occupational lumbar load and lumbar disc herniation as well as lumbar disc narrowing among men and women. Even past lumbar load seems to contribute to the risk of lumbar disc disease. Conclusions According to our study, cumulative physical workload is related to lumbar disc diseases among men and women.
Aim of antiviral therapy of patients with chronic hepatitis C is the sustained elimination of the hepatitis C virus (HCV). The standard of care (SOC) is peginterferon alfa-2a/-2b with ribavirin for 48 weeks or 24 weeks in patients infected with HCV genotype 1 or 2/3, respectively. Overall, approximately half of the patients can be cured by SOC. Based on baseline viral load and the speed of virologic response during treatment, individualization of treatment duration is possible. However, this approach is not sufficient to substantially improve the sustained virologic response (SVR) rates. This goal can be achieved with new HCV specific inhibitors against the NS3/4A polymerase and the NS5B polymerase. Recent trials reported SVR rates in the order of 67-69% and 67-75% for the combination of SOC with the protease inhibitors telaprevir and boceprevir, respectively, in patients with HCV genotype 1 infection. Several new HCV specific inhibitors such as protease inhibitors, nucleoside and non-nucleoside polymerase inhibitors as well as non HCV specific compounds with anti-HCV activity such as cyclophilin inhibitors, silibinin, and nitazoxanide are currently in clinical evaluation. The review describes recent developments and discusses limitations posed by resistance development and drug toxicity.
Although human cytomegalovirus (HCMV) is generally not regarded to be an oncogenic virus, HCMV infection has been implicated in malignant diseases from different cancer entities. On the basis of our experimental findings, we developed the concept of “oncomodulation” to better explain the role of HCMV in cancer. Oncomodulation means that HCMV infects tumor cells and increases their malignancy. By this concept, HCMV was proposed to be a therapeutic target in a fraction of cancer patients. However, the clinical relevance of HCMV-induced oncomodulation remains to be clarified. One central question that has to be definitively answered is if HCMV establishes persistent virus replication in tumor cells or not. In our eyes, recent clinical findings from different groups in glioblastoma patients and especially the detection of a correlation between the numbers of HCMV-infected glioblastoma cells and tumor stage (malignancy) strongly increase the evidence that HCMV may exert oncomodulatory effects. Here, we summarize the currently available knowledge about the molecular mechanisms that may contribute to oncomodulation by HCMV as well as the clinical findings that suggest that a fraction of tumors from different entities is indeed infected with HCMV.
Hepatotoxicity by drugs and dietary supplements (DDS) is a rare and unpredictable event but with the risk of a life-threatening clinical course when it occurs. It may emerge despite intensive chemical, toxicological and observational studies that indicate no hepatotoxic signals. This suggests major clinical and regulatory issues that must be addressed in the area of accurate testing, reporting, and accessibility of reliable data. Consequently, in a clinical setting, safety concerns are key elements in the treatment of patients, and require that the diagnosis of DDS hepatotoxicity clearly be established. Causality of DDS hepatotoxicity may be pursued using a diagnostic algorithm consisting of a pre-test, a main-test as the scale of the updated CIOMS (Council for International Organizations of Medical Sciences), and a post-test. The results of these tests are then sent item by item to the National Health Agency, where the case will undergo further evaluation for pharmacovigilance, strategic aspects and safety issues. After this analysis, all items of the tests are included in the regulatory database freely accessible to the health and scientific community. With this diagnostic and regulatory algorithm the risk of misdiagnoses and inappropriate regulatory measures may be minimized and the safety improved. In conclusion, DDS hepatotoxicity is a rare but is a potentially life-threatening entity requiring a reliable diagnosis with the aid of a diagnostic algorithm, and a thorough pharmacovigilance evaluation by national and international health agencies. Safety aspects in DDS hepatotoxicity represent a major clinical and regulatory issue and should consequently be addressed.
The case of a 64 year old female patient is presented who has treated herself for 9 months with various Indian Ayurvedic herbal products for her vitiligo and experienced a causally related severe hepatotoxicity (ALT, 601 U/L; AST, 663 U/L; Bilirubin, 5.0 mg/dL). After discontinuation, a rapid improvement was observed. Causality assessment with the updated CIOMS (Council for International Organizations of Medical Sciences) scale showed a probable causality (+8 points) for Bakuchi tablets containing extracts from Psoralea corylifolia leaves with psoralens as ingredients, as the primary candidate causing the hepatotoxic reaction. The degree of probability was lower with +6 points for other used herbs: Khadin tablets containing extracts from Acacia catechu leaves; Brahmi tablets containing Eclipta alba or Bacopa monnieri; and Usheer tea prepared from Vetivexia zizaniodis. The case is the first report of Indian Ayurvedic herbal products being potentially hepatotoxic in analogy to some other herbs.
Effects of a phase transition on HBT correlations in an integrated Boltzmann+hydrodynamics approach

A systematic study of HBT radii of pions, produced in heavy ion collisions in the intermediate energy regime (SPS), from an integrated (3+1)d Boltzmann+hydrodynamics approach is presented. The calculations in this hybrid approach, incorporating an hydrodynamic stage into the Ultra-relativistic Quantum Molecular Dynamics transport model, allow for a comparison of different equations of state retaining the same initial conditions and final freeze-out. The results are also compared to the pure cascade transport model calculations in the context of the available data. Furthermore, the effect of different treatments of the hydrodynamic freeze-out procedure on the HBT radii are investigated. It is found that the HBT radii are essentially insensitive to the details of the freeze-out prescription as long as the final hadronic interactions in the cascade are taken into account. The HBT radii RL and RO and the RO/RS ratio are sensitive to the EoS that is employed during the hydrodynamic evolution. We conclude that the increased lifetime in case of a phase transition to a QGP (via a Bag Model equation of state) is not supported by the available data.
The upcoming high energy experiments at the LHC are one of the most outstanding efforts for a better understanding of nature. It is associated with great hopes in the physics community. But there is also some fear in the public, that the conjectured production of mini black holes might lead to a dangerous chain reaction. In this Letter we summarize the most straightforward arguments that are necessary to rule out such doomsday scenarios.
We apply a coupled transport-hydrodynamics model to discuss the production of multi-strange meta-stable objects in Pb + Pb reactions at the FAIR facility. In addition to making predictions for yields of these particles we are able to calculate particle dependent rapidity and momentum distributions. We argue that the FAIR energy regime is the optimal place to search for multi-strange baryonic object (due to the high baryon density, favoring a distillation of strangeness). Additionally, we show results for strangeness and baryon density fluctuations. Using the UrQMD model we calculate the strangeness separation in phase space which might lead to an enhanced production of MEMOs compared to models that assume global thermalization.
The complete molecule of the title compound, C18H24N2O2, is generated by a crystallographic inversion centre. The torsion angles in the hexamethylene chain are consistent with an antiperiplanar conformation, whereas the conformation of the O—CH2—CH2—CH2 unit is gauche. The three-dimensional crystal packing is stabilized by N—H⋯O and N—H⋯N hydrogen bonding.
The title compound, C17H18N2O6, crystallizes with two molecules in the asymmetric unit. In both molecules, one of the C-C bonds of the pentamethylene chain connecting the two aromatic rings is in a trans conformation and another displays a gauche conformation. The aromatic rings within each molecule are nearly coplanar [dihedral angles = 3.36 (9) and 4.50 (9)°] and the nitro groups are twisted slightly out of the planes of their attached rings [dihedral angles = 8.16 (3)/6.6 (2) and 4.9 (4)/3.8 (3)°]. Key indicators: single-crystal X-ray study; T = 173 K; mean σ(C–C) = 0.003 Å; R factor = 0.040; wR factor = 0.101; data-to-parameter ratio = 13.5.
