Acute myeloid leukemia is a hematopoietic stem cell disorder and a type of acute leukemia which is characterized by clonal proliferation of myeloid precursors with a reduced capacity to differentiate into more mature cellular elements. Clinically AML is characterized by a high degree of heterogeneity with respect to chromosome abnormalities, gene mutations, and changes in expression of multiple genes and microRNAs. Cytogenetic abnormalities can be detected in approximately 50% to 60% of newly diagnosed AML patients. Majority of AML cases are associated with chromosomal aberrations, more specifically translocations that often result in gene arrangements and expression of aberrant fusion proteins. This study was carried out with two fusion proteins: PML/RARα and DEK/CAN which results from the translocations t(15;17) and t (6,9) respectively. PML/RARα is the most common translocation (97%) and the main driver in Acute Promyelocytic Leukemia (APL), a wellcharacterized and well treatable subtype of AML. In contrast, DEK/CAN occurs in 1-5% of AML, associated with poor prognosis and defines a high risk group in AML. The expression of PML/RARα results in a fusion protein that acts as a transcriptional repressor by interfering with gene expression programs involved in differentiation, apoptosis, and selfrenewal. Current therapy focused on the targeting of PML/RARα fusion protien. Success has been achieved by using either ATRA, anthracyclines and Arsenic trioxide or their combinations. These agents induce differentiation in PML/RARα positive AML and hence called differentiation therapy. In comparison with ATRA, ATO and anthracyclines are poor cellular differentiation agents. Despite early promise, several studies have reported that differentiation therapy is unable to target/eradicate leukemic stem cells or eradicate the disease. Therefore current therapeutic focus is to eliminate leukemic stem cells and achieve complete molecular remission not only in APL but also in acute lymphoblastic leukemia and chronic myeloid leukemia as well. Key enzymes of the eicosanoid pathways in the arachidonic acid metabolism, such as COX1/2 as well as the 5-LO have been shown to be good targets for leukemic stem cell therapy approach in AML by interfering with the Wntsignaling which is known to be indispensable for the pathogenesis of AML. Recently it was reported that the third eicosanoid pathway based on the cytochrome P450 (CYP) enzymes interferes with Wnt-signaling as well as with the proliferation and mobilization of hematopoietic stem cells...
The aquatic biodiversity of springs and groundwater systems of North Africa remains largely unexplored. In an earlier field survey of Tunisian springs, a new gastropod genus, Bullaregia, was discovered as a phylogenetically independent lineage of uncertain position within the family Hydrobiidae. Here, we provide taxonomic and phylogenetic assignments for three newly collected populations of hydrobiids from springs in northern Tunisia based on morphological, anatomical and genetic (mtCOI and 18S) data. Among these and specimens of Bullaregia, major differences were observed in male and female genitalia as well as in mtCOI sequences (divergence 8.0–9.1%). Based on these findings, we describe two new genera and three new species: Belgrandiellopsis chorfensis gen. et sp. nov., Belgrandiellopsis secunda gen. et sp. nov. and Biserta putealis gen. et sp. nov. In all our phylogenetic analyses, these three new species were well resolved as a monophyletic group together with Bullaregia tunisiensis. Unexpectedly, this clade emerged as sister to the European valvatiform genera Corbellaria and Kerkia and not to the recently discovered clade of groundwater, conchologically similar, species living in Bulgaria (Balkan Peninsula). These Tunisian species are each locally endemic and form part of a newly discovered clade which in future systematic studies could eventually be identified as a distinct hydrobiid subfamily.
The species richness of the aquatic (non-marine) gastropod family Hydrobiidae Stimpson, 1865 reaches its peak in the European region partly because other areas, such as North Africa, remain to be extensively surveyed. Most of the hydrobiid species described in North Africa have been ascribed to the subfamilies Pseudamnicolinae, Hydrobiinae and the genus Mercuria. Little is known about the presence of other hydrobiid subfamilies. This study examines several specimens of gastropods collected from two springs in Tunisia. Based on a comprehensive literature review and rigorous anatomical and molecular comparisons with known species from North Africa, Europe and Asia Minor of similar morphology, we here describe a new genus, Bullaregia gen. nov., and new species Bullaregia tunisiensis gen. et sp. nov. Although the shell shape of this Tunisian species resembles that of European hydrobiid genera such as Belgrandiella, it differs in other anatomical structures (i.e., penis with a glandular strap-like lobe, and two seminal receptacles, SR1 pedunculate, SR2 sessile and less developed). Using maximum likelihood and Bayesian approaches, phylogenetic relationships inferred from mtCOI sequences point to an independent evolutionary lineage for this new taxon outside Belgrandiella and other known hydrobiid subfamilies.
Metabolic critical temperatures define the range of ambient temperatures where endotherms are able to minimize energy allocation to thermogenesis. Examining the relationship between metabolic critical temperatures and basal metabolic rates (BMR) provides a unique opportunity to gain a better understanding of how animals respond to varying ambient climatic conditions, especially in times of ongoing and projected future climate change. We make use of this opportunity by testing the heat dissipation limit (HDL) theory, which hypothesizes that the maximum amount of heat a species can dissipate constrains its energetics. Specifically, we test the theory’s implicit prediction that BMR should be lower under higher metabolic critical temperatures. We analysed the relationship of BMR with upper and lower critical temperatures for a large dataset of 146 endotherm species using regression analyses, carefully accounting for phylogenetic relationships and body mass. We show that metabolic critical temperatures are negatively related with BMR in both birds and mammals. Our results confirm the predictions of the HDL theory, suggesting that metabolic critical temperatures and basal metabolic rates respond in concert to ambient climatic conditions. This implies that heat dissipation capacities of endotherms may be an important factor to take into account in assessments of species’ vulnerability to climate change.
This paper examines the relationship between oil movements and systemic risk of financial institution in major petroleum-based economies. We estimate ΔCoVaR for those institutions and observe the presence of elevated increases in its levels corresponding to the subprime and global financial crises. The results provide evidence in favor of risk measurement improvements by accounting for oil returns in the risk functions. The spread between the standard CoVaR and the CoVaR that includes oil absorbs in a time range longer than the duration of the oil shock. This indicates that the drop in the oil price has a longer effect on risk and requires more time to be discounted by the financial institutions. To support the analysis, we consider also the other major market-based systemic risk measures.
Finding motifs in biological, social, technological, and other types of networks has become a widespread method to gain more knowledge about these networks’ structure and function. However, this task is very computationally demanding, because it is highly associated with the graph isomorphism which is an NP problem (not known to belong to P or NP-complete subsets yet). Accordingly, this research is endeavoring to decrease the need to call NAUTY isomorphism detection method, which is the most time-consuming step in many existing algorithms. The work provides an extremely fast motif detection algorithm called QuateXelero, which has a Quaternary Tree data structure in the heart. The proposed algorithm is based on the well-known ESU (FANMOD) motif detection algorithm. The results of experiments on some standard model networks approve the overal superiority of the proposed algorithm, namely QuateXelero, compared with two of the fastest existing algorithms, G-Tries and Kavosh. QuateXelero is especially fastest in constructing the central data structure of the algorithm from scratch based on the input network.

[MONCl3 · NC - C2Cl3]2 has been prepared by the reaction of MONCl3 with trichloromethyl isocyanidedichloride, CCl3NCCl2 , in CH2Cl2 suspension. The compound forms redbrown. mois-ture sensitive crystals, which were characterized by their IR spectrum as well as by a crystal structure determination (2482 independent observed reflexions, R = 0.048). Crystal data (-70 °C): Space group P21/c, Z = 2, a = 674.2(2); b = 2128.2(11); c = 786.0(4) pm: β = 102.81(3)°. [MONCl3 • NC-C2Cl3]2 forms centrosymmetric dimeric molecules via chloro bridges with Mo-Cl bond lengths of 240.7 and 276.0 pm. The longer MoCl bond of the MOCl2MO ring is caused by the trans influence of the nitride ligand; the MoN bond length of 167 pm corresponds with a triple bond. The 2,3,3-trichloroacrylnitrile ligand is bonded by its nitrogen atom with a bond length of Mo -N = 216 pm; the Mo-N≡C-C sequence is almost linear with a remarkable short C-C bond of 143.0 pm.
This article compares two similar yet never compared cases of intra-European othering: Spain and the South Slavic region. Their common denominator is what I call the Periphery Problem: a hierarchical cultural difference between Europe's symbolic centre (Western Europe) and its exotic peripheries. Using paradigmatic examples intertextually linked to Prosper Mérimée, this article focuses both on the centre (exemplified by Mérimée), and the peripheries' recent responses to Mérimée through meta-images (your image of others' image of you). The structural commonalities in characterization and the entanglements of internal and external images show that national characterization in Europe is profoundly a transnational phenomenon.
Background/Aims: Hepatocellular carcinoma (HCC) represents the second most common cause of cancer-related deaths worldwide, not least due to its high chemoresistance. The long non-coding RNA nuclear paraspeckle assembly transcript 1 (NEAT1), localised in nuclear paraspeckles, has been shown to enhance chemoresistance in several cancer types. Since data on NEAT1 in HCC chemosensitivity are completely lacking and chemoresistance is linked to poor prognosis, we aimed to study NEAT1 expression in HCC chemoresistance and its link to HCC prognosis.
Methods: NEAT1 expression was determined in either sensitive, or sorafenib, or doxorubicin resistant HepG2, PLC/PRF/5, and Huh7 cells by qPCR. Paraspeckles were detected by immunostaining of paraspeckle component 1 (PSPC1) in cell culture and in a cohort of HCC patients. PSPC1 expression was correlated with clinical data. The expression of transcript variants of NEAT1 and transcripts encoding the paraspeckle-associated proteins was analysed in the TCGA liver cancer data set.
Results: NEAT1 was overexpressed in all three sorafenib and doxorubicin resistant cell lines. Paraspeckles were present in all chemoresistant cells, whereas no signal was detected in the sensitive cells. Expression of NEAT1 transcripts as well as transcripts encoding PSPC1, NONO, and RBM14 was increased in tumour tissue. Expression of PSPC1, NONO, and RBM14 transcripts was significantly associated with poor survival, whereas NEAT1 expression was not. Immunohistochemical analysis revealed that nuclear and cytoplasmic PSPC1-positivity was significantly associated with shorter overall survival of HCC patients.
Conclusion: Our data show an induction of NEAT1 in HCC chemoresistance and a high correlation of transcripts encoding paraspeckle-associated proteins with poor survival in HCC. Therefore, NEAT1, PSPC1, NONO, and RBM14 might be promising targets for novel HCC therapies, and the paraspeckle-associated proteins might be clinical markers and predictors for poor survival in HCC.
Reactive oxygen species are a class of naturally occurring, highly reactive molecules that change the structure and function of macromolecules. This can often lead to irreversible intracellular damage. Conversely, they can also cause reversible changes through post-translational modification of proteins which are utilized in the cell for signaling. Most of these modifications occur on specific cysteines. Which structural and physicochemical features contribute to the sensitivity of cysteines to redox modification is currently unclear. Here, I investigated the in uence of protein structural and sequence features on the modifiability of proteins and specific cysteines therein using statistical and machine learning methods. I found several strong structural predictors for redox modification, such as a higher accessibility to the cytosol and a high number of positively charged amino acids in the close vicinity. I detected a high frequency of other post-translational modifications, such as phosphorylation and ubiquitination, near modified cysteines. Distribution of secondary structure elements appears to play a major role in the modifiability of proteins. Utilizing these features, I created models to predict the presence of redox modifiable cysteines in proteins, including human mitochondrial complex I, NKG2E natural killer cell receptors and proximal tubule cell proteins, and compared some of these predictions to earlier experimental results.
Die Dissertation widmet sich der Religionspolitik des oströmischen Kaisers Marcianus, der während der Zeit des Konzils von Chalcedon regierte (450-457). In der Arbeit wird die These aufgestellt, dass der Kaiser einen geradlinigen und konsequenten Kurs in seiner Religionspolitik verfolgte.
Um die These zu untermauern, wurden als Quellen vorwiegend zeitgenössische Dokumente herangezogen: neben Gesetzen und Briefmaterial aus der Feder des Kaisers sowie des römischen Bischofs Leo bestehen diese aus den umfangreichen Konzilsakten.
Die Arbeit zeigt auf, dass sich Marcianus bereits bei seinem Herrschaftsantritt in den dogmatischen Streitigkeiten der Zeit positionierte, indem er sich eindeutig auf die Seite des Dogmas stellte, das der kurz zuvor verstorbene Bischof von Constantinopel Flavianus vertreten hatte, und gleichzeitig das radikal alexandrinische Glaubensbekenntnis ablehnte, das durch den Constantinopolitaner Mönch Eutyches verfochten wurde und dem die Mehrheit seines Reichs im Sommer 450 folgte. Damit setzte er sich von seinem Vorgänger Theodosius II. ab, unter dem es offensichtlich zu keiner Kursänderung in der Religionspolitik gekommen war. Weil das Wohl des Reichs und seine Herrschaft vom orthodoxen Glauben abhingen, war er während seiner gesamten Regierungszeit bemüht, alle für den ‚rechten Glauben‘ zu gewinnen und so die Glaubenseinheit zu wahren. Auch bei den in der Folgezeit des Konzils auftretenden Widerständen gegen die Beschlüsse der Synode setzte Marcianus nicht nur seinen religionspolitischen Kurs durch, sondern verteidigte ziemlich einsam die Glaubensformel des Konzils; selbst in den Widerstandszentren hielt er seinen religionspolitischen Kurs durch, indem er in Palaestina die Aufstände zum Erliegen brachte, in Aegypten mit Hilfe des Militärs den chalcedonischen Bischof zumindest bis zu seinem Tod stützen konnte. Somit kann sein religionspolitischer Kurs von Herrschaftsantritt an bis zu seinem Tod als konsequent und geradlinig bezeichnet werden.
Das World Wide Web (Web) als die wohl wichtigste Entwicklung von Informations- und Kommunikationstechnologien (engl. information and communication technologies, ICTs) Ende des 20. Jahrhunderts bietet uns Zugang zu einer unbegrenzten Fülle an Informationen. Doch um es als reichhaltige Informationsquelle effektiv nutzen zu können, benötigen wir spezifische ICT-Fähigkeiten. Die Dissertation Development of Interactive Performance Measures for two Components of ICT Literacy: Successfully Accessing and Evaluating Information befasst sich daher mit der Untersuchung zweier grundlegender ICT Fähigkeiten für die erfolgreiche Nutzung des Web als Informationsquelle: der Fähigkeit, auf die gewünschten Information zugreifen zu können, also basale Computerfähigkeiten (engl. basic computer skills, BCS), sowie der Fähigkeit, die Online-Informationen in Bezug auf ihre Glaubwürdigkeit bewerten zu können. Hierzu werden zunächst anhand der Betrachtung des theoretischen Hintergrundes beider ICT-Fähigkeiten Definitionen der Konstrukte vorgestellt. Ziel der Arbeit stellt die Entwicklung zweier Testverfahren zur interaktiven, computer-basierten Erfassung basaler Computerfähigkeiten sowie der Fähigkeit zur Bewertung der Glaubwürdigkeit von Online-Informationen dar. Die den Testverfahren zugrundeliegende faktorielle Struktur sowie die Beziehung beider ICT-Fähigkeiten zu verwandten Konstrukten werden untersucht. Des Weiteren werden Ergebnisse aus der praktischen Anwendung des neu entwickelten Tests zur Evaluation von Online-Informationen (TEO) genutzt, um zu einem tieferen Verständnis über den Suchprozess im Web (engl. web search proccess) zu gelangen und Faktoren zu identifizieren, die diesen beeinflussen. Dabei wird sowohl der Einfluss sogenannter Aufgabencharakteristika (engl. task characteristics) untersucht, die den Kontext des Suchprozesses bestimmen, als auch der Einfluss individueller Prozesscharakteristika (engl. individual process characteristics), welche durch die jeweilige Person bestimmt sind, die die Informationssuche vornimmt.
Um den Forschungszielen der Arbeit gerecht zu werden, wurden drei Studien durchgeführt. Die erste Studie befasst sich mit der Entwicklung der Skala zur Erfassung basaler Computerfähigkeiten (BCS). Im Konkreten wurden Hypothesen über die Zusammenhänge der BCS-Skala mit praktischem Computerwissen, Worterkennung, einer Selbsteinschätzung der eigenen Computerfähigkeiten sowie elektronischer Lesefähigkeit formuliert und analysiert. Die zweite Studie behandelt die Entwicklung des Tests zur Evaluation von Online-Informationen (TEO) und exploriert sowohl die latente Struktur des Konstruktes der Bewertung der Glaubwürdigkeit von Online-Informationen als auch den Zusammenhang mit basalen Computerfähigkeiten, Worterkennung und logischem Denken. In der dritten Studie wird der Suchprozess im Web näher beleuchtet und mögliche Einflussgrößen einer erfolgreichen Bewertung von Online-Informationen erforscht, wobei der Einfluss von Aufgabencharakteristika, individuellen Prozesscharakteristika und deren Interaktion ergründet wird. Im Speziellen wurde der Einfluss dreier Aufgabencharakteristika geprüft, die sich auf die Komplexität einer Aufgabe beziehen: die Anzahl der Suchergebnisse (Links auf der Ergebnisseite einer Suchmaschinenabfrage), die Attraktivität der weniger glaubwürdigen Links auf der Ergebnisseite einer Suchmaschinenabfrage im Vergleich zum glaubwürdigsten Link sowie die Kongruenz zwischen den Glaubwürdigkeitsmerkmalen in den Links auf der Ergebnisseite der Suchmaschinenabfrage und auf den dazugehörigen Webseiten. Dabei war die Attraktivität eines Links definiert als die Gesamtanzahl der Glaubwürdigkeitsmerkmale, welche auf eine hohe Glaubwürdigkeit der Informationen hinweisen. Je mehr Merkmale eines Links hohe Glaubwürdigkeit indizieren, desto attraktiver ist ein Link. Von Kongruenz zwischen den Glaubwürdigkeitsmerkmalen wurde ausgegangen, wenn diese sowohl in einem Link auf der Ergebnisseite der Suchmaschinenabfrage als auch auf der korrespondierenden Webseite gleichermaßen hohe oder gleichermaßen geringe Glaubwürdigkeit anzeigen. Inkongruenz hingegen wurde angenommen, wenn die Glaubwürdigkeitsmerkmale in einem Link auf der Ergebnisseite hohe Glaubwürdigkeit und jene auf der korrespondierenden Webseite geringe Glaubwürdigkeit anzeigen und vice versa. Weiterhin wurde der Einfluss dreier individueller Prozesscharakteristika auf den Bewertungserfolg untersucht: die Anzahl besuchter unterschiedlicher Webseiten, die auf der Ergebnisseite der Suchmaschinenabfrage verbrachte Zeit sowie die auf den korrespondierenden Webseiten verbrachte Zeit.
Zusammengefasst präsentiert die Arbeit reliable Instrumente zur Erfassung basaler Computerfähigkeiten sowie der Fähigkeit zur Bewertung der Glaubwürdigkeit von Online-Informationen. Sie zeigt die hohe Relevanz basaler Lesefähigkeiten (Worterkennung) für beide Konstrukte auf und offenbart die Fähigkeit zum logischen Denken als Prädiktor für die Bewertungskompetenz. Während eine erfolgreiche Bewertung von Online-Informationen durch eine hohe Anzahl von Suchergebnissen negativ beeinflusst wurde, wirkte sich Kongruenz der Glaubwürdigkeitsmerkmale sowie eine hohe Anzahl besuchter unterschiedlicher Webseiten positiv auf den Bewertungserfolg aus. Die Attraktivität der weniger glaubwürdigen Links sowie die zeitbezogenen Prozesscharakteristika beeinflussten den Bewertungserfolg wider Erwarten nicht. Die Anzahl besuchter unterschiedlicher Webseiten erwies sich bei einer hohen Anzahl von Suchergebnissen als weniger prädiktiv für den Bewertungserfolg als bei einer geringen Anzahl von Suchergebnissen.
Iron deprivation activates mitophagy and extends lifespan in nematodes. In patients suffering from Parkinson’s disease (PD), PINK1-PRKN mutations via deficient mitophagy trigger iron accumulation and reduce lifespan. To evaluate molecular effects of iron chelator drugs as a potential PD therapy, we assessed fibroblasts by global proteome profiles and targeted transcript analyses. In mouse cells, iron shortage decreased protein abundance for iron-binding nucleotide metabolism enzymes (prominently XDH and ferritin homolog RRM2). It also decreased the expression of factors with a role for nucleotide surveillance, which associate with iron-sulfur-clusters (ISC), and are important for growth and survival. This widespread effect included prominently Nthl1-Ppat-Bdh2, but also mitochondrial Glrx5-Nfu1-Bola1, cytosolic Aco1-Abce1-Tyw5, and nuclear Dna2-Elp3-Pold1-Prim2. Incidentally, upregulated Pink1-Prkn levels explained mitophagy induction, the downregulated expression of Slc25a28 suggested it to function in iron export. The impact of PINK1 mutations in mouse and patient cells was pronounced only after iron overload, causing hyperreactive expression of ribosomal surveillance factor Abce1 and of ferritin, despite ferritin translation being repressed by IRP1. This misregulation might be explained by the deficiency of the ISC-biogenesis factor GLRX5. Our systematic survey suggests mitochondrial ISC-biogenesis and post-transcriptional iron regulation to be important in the decision, whether organisms undergo PD pathogenesis or healthy aging.
Biallelic pathogenic variants in CLPP, encoding mitochondrial matrix peptidase ClpP, cause a rare autosomal recessive condition, Perrault syndrome type 3 (PRLTS3). It is characterized by primary ovarian insufficiency and early sensorineural hearing loss, often associated with progressive neurological deficits. Mouse models showed that accumulations of (i) its main protein interactor, the substrate-selecting AAA+ ATPase ClpX, (ii) mitoribosomes, and (iii) mtDNA nucleoids are the main cellular consequences of ClpP absence. However, the sequence of these events and their validity in human remain unclear. Here, we studied global proteome profiles to define ClpP substrates among mitochondrial ClpX interactors, which accumulated consistently in ClpP-null mouse embryonal fibroblasts and brains. Validation work included novel ClpP-mutant patient fibroblast proteomics. ClpX co-accumulated in mitochondria with the nucleoid component POLDIP2, the mitochondrial poly(A) mRNA granule element LRPPRC, and tRNA processing factor GFM1 (in mouse, also GRSF1). Only in mouse did accumulated ClpX, GFM1, and GRSF1 appear in nuclear fractions. Mitoribosomal accumulation was minor. Consistent accumulations in murine and human fibroblasts also affected multimerizing factors not known as ClpX interactors, namely, OAT, ASS1, ACADVL, STOM, PRDX3, PC, MUT, ALDH2, PMPCB, UQCRC2, and ACADSB, but the impact on downstream metabolites was marginal. Our data demonstrate the primary impact of ClpXP on the assembly of proteins with nucleic acids and show nucleoid enlargement in human as a key consequence.
Human RNF213, which encodes the protein mysterin, is a known susceptibility gene for moyamoya disease (MMD), a cerebrovascular condition with occlusive lesions and compensatory angiogenesis. Mysterin mutations, together with exposure to environmental trigger factors, lead to an elevated stroke risk since childhood. Mysterin is induced during cell stress, to function as cytosolic AAA+ ATPase and ubiquitylation enzyme. Little knowledge exists, in which context mysterin is needed. Here, we found that genetic ablation of several mitochondrial matrix factors, such as the peptidase ClpP, the transcription factor Tfam, as well as the peptidase and AAA+ ATPase Lonp1, potently induces Rnf213 transcript expression in various organs, in parallel with other components of the innate immune system. Mostly in mouse fibroblasts and human endothelial cells, the Rnf213 levels showed prominent upregulation upon Poly(I:C)-triggered TLR3-mediated responses to dsRNA toxicity, as well as upon interferon gamma treatment. Only partial suppression of Rnf213 induction was achieved by C16 as an antagonist of PKR (dsRNA-dependent protein kinase). Since dysfunctional mitochondria were recently reported to release immune-stimulatory dsRNA into the cytosol, our results suggest that mysterin becomes relevant when mitochondrial dysfunction or infections have triggered RNA-dependent inflammation. Thus, MMD has similarities with vasculopathies that involve altered nucleotide processing, such as Aicardi-Goutières syndrome or systemic lupus erythematosus. Furthermore, in MMD, the low penetrance of RNF213 mutations might be modified by dysfunctions in mitochondria or the TLR3 pathway.
Depletion of yeast/fly Ataxin-2 rescues TDP-43 overexpression toxicity. In mouse models of Amyotrophic Lateral Sclerosis via TDP-43 overexpression, depletion of its ortholog ATXN2 mitigated motor neuron degeneration and extended lifespan from 25 days to >300 days. There is another ortholog in mammals, named ATXN2L (Ataxin-2-like), which is almost uncharacterized but also functions in RNA surveillance at stress granules. We generated mice with Crispr/Cas9-mediated deletion of Atxn2l exons 5-8, studying homozygotes prenatally and heterozygotes during aging. Our novel findings indicate that ATXN2L absence triggers mid-gestational embryonic lethality, affecting female animals more strongly. Weight and development stages of homozygous mutants were reduced. Placenta phenotypes were not apparent, but brain histology showed lamination defects and apoptosis. Aged heterozygotes showed no locomotor deficits or weight loss over 12 months. Null mutants in vivo displayed compensatory efforts to maximize Atxn2l expression, which were prevented upon nutrient abundance in vitro. Mouse embryonal fibroblast cells revealed more multinucleated giant cells upon ATXN2L deficiency. In addition, in human neural cells, transcript levels of ATXN2L were induced upon starvation and glucose and amino acids exposure, but this induction was partially prevented by serum or low cholesterol administration. Neither ATXN2L depletion triggered dysregulation of ATXN2, nor a converse effect was observed. Overall, this essential role of ATXN2L for embryogenesis raises questions about its role in neurodegenerative diseases and neuroprotective therapies.

Here, avoiding recombinant overexpression, we employed complexomics in mitochondria from three mouse tissues to identify endogenous targets. CLPP absence caused accumulation and dispersion of CLPX-VWA8 as AAA+ unfoldases, and of PLPBP. Similar changes and CLPX-VWA8 comigration were evident for mitoribosomal central protuberance clusters, translation factors like GFM1-HARS2, RNA granule components LRPPRC-SLIRP, and enzymes OAT-ALDH18A1. Mitochondrially translated proteins in testis showed reductions to <30% for MTCO1-3, misassembly of complex-IV supercomplex, and accumulated metal-binding assembly factors COX15-SFXN4. Indeed, heavy metal levels were increased for iron, molybdenum, cobalt and manganese. RT-qPCR showed compensatory downregulation only for Clpx mRNA, most accumulated proteins appeared transcriptionally upregulated. Immunoblots validated VWA8, MRPL38, MRPL18, GFM1 and OAT accumulation. Coimmunoprecipitation confirmed CLPX binding to MRPL38, GFM1 and OAT, so excess CLPX and PLP may affect their activity. Our data elucidate mechanistically the mitochondrial translation fidelity deficits, which underlie progressive hearing impairment in PRLTS3.

Morbus Parkinson (abgekürzt als PD vom Englischen Parkinson’s disease) ist nach Alzheimer die zweithäufigste neurodegenerative Erkrankung. Die Hauptmerkmale sind Rigidität und Bradykinesie, sowie Tremor und posturale Instabilität. Im Gehirn lässt sich bei Parkinsonpatienten post mortem ein Verlust an Neuronen in der Substantia nigra feststellen, was zu den ersten beiden Anzeichen führt. Zudem gibt es intrazelluläre Einschlüsse in den betroffenen Nervenzellen – Lewy-Körperchen genannt – die aus Alpha-Synuklein und anderen Proteinen wie Ubiquitin zusammengesetzt sind. Außerdem ist der Eisenmetabolismus in Gehirnen von Parkinsonpatienten gestört und man findet Eisen-Ablagerungen, vor allem im Mittelhirn. Die Ursachen für PD sind bislang nicht abschließend geklärt. Der Großteil der Fälle ist sporadischer Natur mit unbekannter Ursache und nur bei einem geringen Anteil liegt eine Mutation in einem einzelnen Gen zugrunde. Die häufigsten Mutationen tritt in den Genen für Alpha-Synuklein (SNCA), PINK1 und PARKIN auf.
Die Serin-Threonin-Kinase PINK1 und die E3-Ubiquitin-Protein-Ligase PARKIN sind zwei Proteine, die in Stresssituationen an der Mitochondrien-Außenmembran am Abbau von alten oder nicht richtig funktionierenden Mitochondrien beteiligt sind. Dieser Vorgang nennt sich Mitophagie.
Die dieser Arbeit zugrunde liegenden Publikationen gehen den Zusammenhängen zwischen mitochondrialen Fehlfunktionen und der Pathogenese von PD nach. Da die Krankheit meist erst im hohen Alter auftritt, davon größtenteils ohne direkte Ursache, liegt der Schluss nahe, dass neben genetischen Ursachen auch Umweltfaktoren eine größere Rolle spielen könnten. Um dies näher zu analysieren, wurden experimentell verschiedene Stressoren eingesetzt.
Insgesamt wurden folgende Aspekte untersucht:
I. Welche Auswirkungen hat das Fehlen von PINK1 auf die Zelle? Gibt es einen Biomarker, der mit höherem Alter immer stärker verändert ist?
II. Welchen Einfluss haben Umweltfaktoren wie veränderte Eisen-Exposition auf die Zelle und was verändert sich beim Fehlen von PINK1?
III. Wie können mitochondriale Fehlfunktionen präferentiell das Nervensystem betreffen, wenn es nicht um respiratorische Insuffizienz geht?
Die einzelnen Studien zeigten folgende Ergebnisse:
Torres-Odio/Key et al. 2017 widmete sich der Suche nach molekularen Biomarkern, wodurch PD präsymptomatisch erkannt und die Progression der Erkrankung eingeschätzt werden kann. Die Transkriptom-Analyse der Kleinhirne von Mäusen mit Pink1-/--Mutation in drei verschiedenen Altersstufen zeigte eindrücklich, dass nicht ein einzelner Faktor immer stärker verändert war, sondern, dass immer mehr Faktoren und daher auch eine steigende Zahl an
Signalwegen mit höherem Alter beteiligt waren. Diese Veränderungen betrafen inflammatorische Signalwege, insbesondere Faktoren, die mit der Erkennung und Verarbeitung von zellfremden Nukleinsäuren assoziiert sind. Aufgrund der evolutionären Herkunft von Mitochondrien als frühere Protobakterien haben mitochondriale Nukleinsäuren und Proteine zum Teil bakterielle Ähnlichkeiten, und könnten bei Fehlfunktionen ins Zytosol gelangen. Vor diesem Hintergrund lassen die Ergebnisse der Studie den Schluss zu, dass das angeborene Immunsystem in Neuronen durch eine PINK1-assoziierte mitochondriale Störung aktiviert wird.
In der Publikation Key et al. 2020 wurde Eisen als ein im täglichen Leben vorkommender Stressor eingesetzt und es wurden systematisch Faktoren des Eisenstoffwechsels bei hohen und niedrigen Eisenspiegeln im Zusammenhang mit Parkinson-Mutationen untersucht. Da Eisen für die Gesundheit von Mitochondrien eine große Rolle spielt und Eisen-Chelatoren als Therapie bei PD Patienten bereits diskutiert werden, haben die molekularen Befunde große Relevanz. Die Ergebnisse zeigen, dass unter niedrigen Eisenspiegeln Proteine reduziert waren, die am Nukleotid-Stoffwechsel beteiligt sind, sowie Faktoren, die Eisen-Schwefel-Cluster als Cofaktoren haben und wichtig für die Nukleotid-Qualitätskontrolle sind. Das Fehlen von Eisen führte zu einer Induktion von Pink1 und Prkn, was auf verstärkte Mitophagie hindeutet. Insgesamt konnte gezeigt werden, dass die mitochondriale Eisen-Schwefel-Cluster Biogenese und die post-transkriptionelle Eisenregulation entscheidend für die Pathogenese von PD, bzw. das gesunde Fortbestehen einer Zelle und letztlich auch eines Organismus sind.
In Key et al. 2019 wurde erstmalig das Gesamt-Ubiquitylom aus Gehirnen von gealterten Parkin-knockout (KO) Mäusen erhoben und analysiert, um Ubiquitylierungs-Substrate von PARKIN zu identifizieren. Hierbei zeigte sich eine veränderte Ubiquitylierung von mehreren Faktoren, die an der zellulären Calcium-Homöostase beteiligt sind. Weitere elektrophysiologische Experimente in Gehirnen von gealterten Parkin-/--KO Mäusen ergaben, dass in Nervenzellen im Locus coeruleus die Geschwindigkeit der spontanen Taktgeber erhöht, dass die langsame Nachhyperpolarisation reduziert und, dass die Dauer der Aktionspotentiale erniedrigt war, ohne Veränderung der Kaliumkanal-Ströme.
Insgesamt geht aus den drei Studien hervor, dass mitochondriale Fehlfunktionen bei dauerhaftem Bestehen weitreichende Folgen für die Gesundheit des Nervensystems haben können, denn auch kleine Veränderungen, seien es durch Mutationen oder Umweltfaktoren wie Eisen, können in einer so großen Lebensspanne wie der des Menschen über Krankheit oder Gesundheit entscheiden!
This article demonstrates the use of guided elastic waves (GEW) for multiple-in and multiple-out (MIMO) data communication in the framework of a structural health monitoring (SHM) system. Therefore, miniaturized low-voltage communication nodes have been developed. They are arranged in a spatially distributed and permanently installed network. Wireless exchange of encoded information across a metallic plate and a stiffened carbon-fiber reinforced plastics (CFRP) structure is investigated. A combination of square-wave excitation sequences and frequency-division multiplexing (FDM) is explored for parallel communication with multiple nodes. Moreover, the impact of the excitation-sequence length on the reliability of information transmission is studied in view of future energy-aware application scenarios. The presented system achieves in both studied structures error-free transmission at a data rate of 0.17 kbps (per carrier frequency) with a power consumption of 224 mW.
Capturing intermolecular interactions accurately is essential for describing, e.g., morphology of molecular matter on the nanoscale. When it reveals characteristics which are not directly accessible through experiments or ab initio theories, a model here becomes eminently beneficial. In laboratory astrochemistry, the intense study of ices has led i.a. to the exploration of the spontelectric state of nanofilms. Despite its success in biophysics or biochemistry and despite its predictive power, molecular modeling has however not yet been widely deployed for solid-state astrochemistry. In this article, therefore a pertinent hitherto unaddressed problem is tackled by means of the classical molecular-dynamics method, namely the unknown distribution of relative dipole orientations in spontelectric cis-methyl formate (MF). In doing so, from ab initio data, a molecular model is derived which confirms for the first time the anomalous temperature-dependent polarization of MF. These insights thus represent a further step toward understanding spontelectric behavior. Moreover, unprecedented first-principles predictions are reported regarding the ground-state geometry of the MF trimer and tetramer. In conjunction with the study of the binding to carbonaceous substrates, these additional findings can help to exemplarily elucidate molecular ice formation in astrochemical settings.
Sparse sensor networks for Lamb wave-based structural health monitoring (SHM) can detect defects in plate-like structures. However, the limited number of sensor positions provides little information to characterize the unknown scatterer. This can be achieved by full wavefield analysis e.g. using Laser Doppler vibrometry measurements.
This paper proposes deconvolution processing that enhances the acoustic wavefield interpretation by increasing the temporal resolution of the underlying ultrasound signals. Applying this preprocessor to the whole wavefield allows improved non-destructive assessment of the defect. This approach is verified experimentally through a case study on an isotropic aluminum plate with four cracks.
Internet Gaming Disorder (IGD) has been included in the DSM-5 as a diagnosis for further study, and Gaming Disorder as a new diagnosis in the ICD-11. Nonetheless, little is known about the clinical prevalence of IGD in children and adolescents. Additionally, it is unclear if patients with IGD are already identified in routine psychotherapy, using the ICD-10 diagnosis F 63.8 (recommended classification of IGD in ICD-10). This study investigated N = 358 children and adolescents (self and parental rating) of an outpatient psychotherapy centre in Germany using the Video Game Dependency Scale. According to self-report 4.0% of the 11- to 17-year-old patients met criteria for a tentative IGD diagnosis and 14.0% according to the parental report. Of the 5- to 10-year-old patients, 4.1% were diagnosed with tentative IGD according to parental report. Patients meeting IGD criteria were most frequently diagnosed with hyperkinetic disorders, followed by anxiety disorders, F 63.8, conduct disorders, mood disorders and obsessive-compulsive disorders (descending order) as primary clinical diagnoses. Consequently, this study indicates that a significant amount of the clinical population presents IGD. Meaning, appropriate diagnostics should be included in routine psychological diagnostics in order to avoid “hidden” cases of IGD in the future.
During the last decade, three new acidophilous forests associations were detected in the Mecsek Mts (SW Hungary), and described as acidophilous beech wood (Sorbo torminalis-Fagetum (A. O. Horvat 1963a) Borhidi et Kevey in Kevey 2001), acido-mesophilous oak wood (Luzulo forsteri-Quercetum petraeae (A. O. Horvat 1963a) Borhidi et Kevey 1996) and acido-xerophilous oak shrubland (Genisto pilosae-Quercetum polycarpae (A. O. Horvat 1967) Borhidi et Kevey 1996). In this article two further new associations are described: the acidophilous oakwood of the Mecsek (Viscario-Quercetum polycarpae Kevey, ass. nova) and the acido-mesophilous oakwood of western Hungary (Campanulo rotundifoliae-Quercetum petraeae (Csapody 1964) Kevey, ass. nova). These associations are related to the acidophilous forests of the Balkan Peninsula based on the infrequent presence of sub-Mediterranean species. A detailed comparative study of these new associations with the earlier known ones permitted to develop a reshaped classification of the syntaxonomy of these units, creating four new suballiances: within the frame of Quercion farnetto I. Horvat 1938 the suballiances Luzulo forsteri-Quercenion polycarpae Kevey, suball. nova and the typical Quercenion farnetto Kevey, suball. nova, in the frame of Quercion petraeae Zolyomi et Jakucs 1957 the suballiances Luzulo multiflorae-Quercenion petraeae Kevey, suball. nova and the Quercenion petraeae Kevey, suball. nova.
Background: Mastectomy in male transgender patients is an important (and often the first) step toward physical manhood. At our department, mastectomies in transgender patients have been performed for several decades.
Methods: Recorded data were collected and analyzed for all male transgender patients undergoing mastectomy over a period of 24 years at our department.
Results: In total, 268 gender-reassigning mastectomies were performed. Several different mastectomy techniques (areolar incision, n=172; sub-mammary incision, n=96) were used according to patients' habitus and breast features. Corresponding to algorithms presented in the current literature, certain breast qualities were matched with a particular mastectomy technique. Overall, small breasts with marginal ptosis and good skin elasticity allowed small areolar incisions as a method of access for glandular removal. In contrast, large breasts and those with heavy ptosis or poor skin elasticity often required larger incisions for breast amputation. The secondary correction rate (38%) was high for gender reassignment mastectomy, as is also reflected by data in the current literature. Secondary correction frequently involved revision of chest wall recontouring, suggesting inadequate removal of the mammary tissue, as well as scar revision, which may reflect intense traction during wound healing (36%). Secondary corrections were performed more often after using small areolar incision techniques (48%) than after using large sub-mammary incisions (21%).
Conclusions: Choosing the suitable mastectomy technique for each patient requires careful individual evaluation of breast features such as size, degree of ptosis, and skin elasticity in order to maximize patient satisfaction and minimize secondary revisions.
A feature of the Northern Iroquoian languages is their especially rich inventory of particles. This paper is concerned with one particle in the Cayuga language which has a widespread distribution and performs a broad range of apparently unrelated functions. The particle ne:' is commonly .translated as 'it is/that is', 'this' or ' that'. In other instances it is translated as predominant stress, or is simply omitted in the translation. The particle can occur in almost any syntactic or semantic environment, but it is not obligatory in any context. The various functions that have been suggested in the literature include indication of declarative mood and assertion, marking of emphasis, focus or contrast, and expression of predicative and deictic force. I argue that the particle ne:' can be described successfully if its distribution is considered from a wider perspective, taking into account discourse structure and variation in scope. Its analysis as a focus marker can account for the variety of apparently unrelated functions. The analysis is based on a detailed study of the particle' s distribution in spoken language using a database of five Cayuga texts by four different speakers, including three narratives, one procedural text and a children 's version of a ceremonial text.
Am Peischlachtörl (2484 m Meereshöhe, nördliche Schobergruppe, Nationalpark Hohe Tauern) wurden Bültenböden vegetations- und bodenkundlich untersucht und mit einem typischen alpinen Rasen (Caricetum curvulae) verglichen. Insgesamt wurden 380 Vegetationsaufnahmen à 10 x 10 cm entlang von 14 Kleintransekten durchgeführt, um das Vegetationsmosaik von Bülten und Senken darzustellen. Die Untersuchungen umfassten auch die Bestimmung der Bodentypen, pH-Messungen und Korngrößenanalysen. Bodentemperaturmessungen erfolgten in 10 und 50 cm Bodentiefe von 2003 bis 2005. Es zeigte sich eine deutliche Vegetationszonierung von den Bülten zu den Senken. Die Bultkuppen werden von einer Gesellschaft bestimmt, die dem Caricetum curvulae zugeordnet werden kann, mit einer Loiseleuria procumbens-Ausbildung an einer kleineren Anzahl von Kuppen, einer Oreochloa disticha-Ausbildung für den überwiegenden Anteil der Kuppen sowie einer Ligusticum mutellina-Ausbildung im Übergang zu den Senken. Der alpine Rasen (Caricetum curvulae) lässt sich gut mit der Vegetation der Bultkuppen vergleichen, wobei der überwiegende Teil der Rasen-Aufnahmen zur Oreochloa disticha-Ausbildung gezählt werden kann. Die Senken der Bültenböden werden von einer Luzula alpinopilosa-Ausbildung charakterisiert. Eine pflanzensoziologische Zuordnung dieser Gesellschaft ist sehr schwierig, da Schneebodenarten, Gräser der subalpinen Stufe und Caricetum curvulae-Arten vermischt vorkommen. Im Bereich der Bülten wurde eine pseudovergleyte Braunerde bzw. ein Semipodsol festgestellt, in der Senke ein Pseudogley und im alpinen Rasen ein Eisen-Humus-Podsol. Bültenböden und alpiner Rasen unterscheiden sich signifikant hinsichtlich der Korngrößen: der Boden des alpinen Rasens ist wesentlich skelett- und sandreicher. Kuppen und Senken unterscheiden sich nur unwesentlich in ihrer Körnung. - Der erste Winter zeichnete sich durch fehlenden Bodenfrost aus, während im zweiten Winter sowohl der Boden des alpinen Rasens als auch die Bülten tief durchfroren. Die Temperatur in den Senken sank dagegen nur leicht unter 0 °C ab.
Using hand-collected data on CEO appointments during shareholder activism campaigns, this study examines whether shareholder involvement in CEO recruiting affects frictions in CEO hiring decisions. The results indicate that appointments of CEOs who are recruited with shareholder activist influence are followed by more favorable stock market reactions and stronger profitability improvements than CEO appointments that also occur during activism campaigns but without the influence of activists. I find little evidence that shareholder activists increase hiring frictions by facilitating the recruiting of CEOs who will implement myopic corporate policies. Analyses of recruiting process characteristics reveal that activist influence is associated with more resources being dedicated to the CEO search process and with a higher propensity to recruit CEOs from outside the firm. These findings contribute to the CEO labor market literature, which tends to focus on the decision to remove incumbent CEOs but provides limited insights into CEO recruiting.
In this contribution, I would like to examine the way in which Bakhtin, in the two essays dedicated to the chronotope, lays the foundations for a theory of literary imagination. […] His concept of the chronotope may be interpreted as a contribution to a tradition in which Henri Bergson, William James, Charles Sander Peirce and Gilles Deleuze have been key figures. Like these four authors, Bakhtin is a philosopher in the school of pragmatism. His predilection for what Gary Saul Morson and Caryl Emerson have called “prosaics” puts him right at the heart of a philosophical family that calls forth multiplicity against metaphysical essentialism, and prefers the mundane to the universal. It seems wise to proceed carefully in the attempt to reconstruct Bakhtin’s theory of imagination. In this contribution to the debate, I choose to develop a philosophical dialogue between Bakhtin and the above-mentioned philosophical family. More specifically, it seems to me that the ideal point of departure for examining the way in which Bakhtin attempts to get to the bottom of the mysteries of literary imagination is Gilles Deleuze’s synthesis of Bergson’s epistemological view on knowledge as “the perception of images”, as well as Peirce’s theory of experience based on a typology of images. In the following, I show that Bakhtin’s view of the temporal-spatial constellations in literature demonstrates a strong affinity to the Bergsonian view that perception of the spatial world is colored by the lived time experienced by the observer. Based on this observation, I then develop a typology of images which places the concept of the chronotope in a more systematic framework.

Mongolian spots (MS) are congenital dermal conditions resulting from neural crest-derived melanocytes migration to the skin during embryogenesis. MS incidences are highly variable in different populations. Morphologically, MS present as hyperpigmented maculae of varying size and form, ranging from round spots of 1 cm in diameter to extensive discolorations covering predominantly the lower back and buttocks. Due to their coloring, which is also dependent on the skin type, MS may mimic hematoma thus posing a challenge on the physician conducting examinations of children in cases of suspected child abuse. In the present study, MS incidences and distribution, as well as skin types, were documented in a collective of 253 children examined on the basis of suspected child abuse. From these data, a classification scheme was derived to document MS and to help identify cases with a need for recurrent examination for unambiguous interpretation of initial findings alongside the main decisive factors for re-examination such as general circumstances of the initial examination (e. g., experience of the examiner, lighting conditions) and given dermatological conditions of the patient (e. g., diaper rash).
Der Beitrag versucht die Lektüre einer wenig bekannten Erzählung Oskar Panizzas unter machttheoretischen Aspekten. Mit ihrer Fokussierung auf Panizzas Kritik an der Hegemonie des wilhelminischen Kaiserreiches reproduziert die Panizza-Forschung ein Machtverständnis, das Macht als Besitz der Herrschenden konzipiert. Ein solches Verständnis verstellt den Blick auf die produktive Qualität und die performative Wirkung von Panizzas Texten. In Anlehnung an Michel Foucaults Konzept von Macht als produktivem Netz von Kräfteverhältnissen zeigt mein Beitrag, wie sich das Verhältnis zwischen Norm und Abweichung nicht in der zentralen Ausübung einer einzelnen Machtinstanz, sondern in dem dezentralen und produktiven Spiel der Kräfteverhältnisse beschreiben lässt. Vermittels literarischer Techniken inszeniert Panizzas Erzählung Ein scandalöser Fall ein solch dynamisches Machtverständnis und macht dabei die prekäre Beschaffenheit hegemonialer Konstellationen sichtbar.
Aim: To evaluate the long-term outcome of catheter ablation of atrial fibrillation (AF) facilitated by preprocedural three-dimensional (3-D) transesophageal echocardiography.
Methods: In 50 patients, 3D transesophageal echocardiography (3D TEE) was performed immediately prior to an ablation procedure (paroxysmal AF: 30 patients, persistent AF: 20 patients). The images were available throughout the ablation procedure. Two different ablation strategies were used. In most of the patients with paroxysmal AF, the cryoablation technique was used (Arctic Front Balloon, CryoCath Technologies/Medtronic; group A2). In the other patients, a circumferential pulmonary vein ablation was performed using the CARTO system [Biosense Webster; group A1 (paroxysmal AF), group B (persistent AF)]. Success rates and complication rates were analysed at 4-year follow-up.
Results: A 3D TEE could be performed successfully in all patients prior to the ablation procedure and all four pulmonary vein ostia could be evaluated in 84% of patients. The image quality was excellent in the majority of patients and several variations of the pulmonary vein anatomy could be visualized precisely (e.g., common pulmonary vein ostia, accessory pulmonary veins, varying diameter of the left atrial appendage and its distance to the left superior pulmonary vein). All ablation procedures could be performed as planned and almost all pulmonary veins could be isolated successfully. At 48-mo follow-up, 68.0% of all patients were free from an arrhythmia recurrence (group A1: 72.7%, group A2: 73.7%, group B: 60.0%). There were no major complications.
Conclusion: 3D TEE provides an excellent overview over the left atrial anatomy prior to AF ablation procedures and these procedures are associated with a favourable long-term outcome.
Aim: To evaluate the effectiveness of two different strategies using radiofrequency catheter ablation for redo procedures after cryoablation of atrial fibrillation.
Methods: Thirty patients (paroxysmal atrial fibrillation: 22 patients, persistent atrial fibrillation: 8 patients) had to undergo a redo procedure after initially successful circumferential pulmonary vein (PV) isolation with the cryoballoon technique (Arctic Front Balloon, CryoCath Technologies/Medtronic). The redo ablation procedures were performed using a segmental approach or a circumferential ablation strategy (CARTO; Biosense Webster) depending on the intra-procedural findings. After discharge, patients were scheduled for repeated visits at the arrhythmia clinic. A 7-day Holter monitoring was performed at 3, 12 and 24 mo after the ablation procedure.
Results: During the redo procedure, a mean number of 2.9 re-conducting pulmonary veins (SD ± 1.0 PVs) were detected (using a circular mapping catheter). In 20 patients, a segmental approach was sufficient to eliminate the residual pulmonary vein conduction because there were only a few recovered pulmonary vein fibres. In the remaining 10 patients, a circumferential ablation strategy was used because of a complete recovery of the PV-LA conduction. All recovered pulmonary veins could be isolated successfully again. At 2-year follow-up, 73.3% of all patients were free from an arrhythmia recurrence (22/30). There were no major complications.
Conclusion: In patients with an initial circumferential pulmonary vein isolation using the cryoballoon technique, a repeat ablation procedure can be performed safely and effectively using radiofrequency catheter ablation.
In situ investigation of membrane proteins is a challenging task. Previously we demonstrated that nitroxide labels combined with pulsed ESR spectroscopy is a promising tool for this purpose. However, the nitroxide labels suffer from poor stability, high background labeling, and low sensitivity. Here we show that Finland (FTAM) and OX063 based labels enable labeling of the cobalamin transporter BtuB and BamA, the central component of the β-barrel assembly machinery (BAM) complex, in E coli. Compared to the methanethiosulfonate spin label (MTSL), trityl labels eliminated the background signals and enabled specific in situ labeling of the proteins with high efficiency. The OX063 labels show a long phase memory time (TM) of ≈5 μs. All the trityls enabled distance measurements between BtuB and an orthogonally labeled substrate with high selectivity and sensitivity down to a few μm concentration. Our data corroborate the BtuB and BamA conformations in the cellular environment of E. coli.

In der hier zu besprechenden Arbeit von R. Brendel, einer überarbeiteten und ergänzten Fassung der an der LMU München im Oktober 2013 eingereichten Dissertation, werden, wie es der Verlag Dr. Kovač ankündigt, "erstmals vollständig sämtliche mit Julian in Verbindung stehende[n] Gesetzestexte gesammelt, in Übersetzung vorgelegt und ausgewertet". Neben den Werken von S. Conti über die Inschriften Julians und Th. Fleck über die Portraits wird damit ein weiteres Themenfeld für den Kaiser systematisch erschlossen, der nach wie vor moderne Forscher in seinen Bann zieht. Als methodisch weiterbringend erachtet der Verf. es auch, die spätantiken und frühmittelalterlichen Kommentare wie die Summaria antiqua Codicis Theodosiani und die Summa Perusina Codicis Iustiniani (zit. S. 446) heranzuziehen. ...
Die von J. Wiesehöfer herausgegebene Reihe Oriens et Occidens ist um einen neuen, wissenschaftsgeschichtlich wie kulturhistorisch wertvollen Band bereichert worden, den inzwischen 22. der 2001 begonnenen Reihe. Verfasst hat ihn der emeritierte amerikanische Gelehrte Lawrence J. Baack (* 1943), der sich u.a. durch seine Forschungen zur deutschen Geschichte des frühen 19. Jhs. einen Namen gemacht hat und auch über dänische und schwedische Sprachkenntnisse verfügt. Schon der fünfte Band der Reihe war derselben Thematik gewidmet. Er ist als Begleitband höchst wertvoll mit seinen zahlreichen Spezialbeiträgen, die in dieser Ausführlichkeit vom Vf. nicht behandelt werden konnten. Der Titel darf – auch wenn in der Forschung oft von der „Arabischen Reise“ gesprochen wird – nicht darüber hinwegtäuschen, dass zwar das Hauptziel der von J. D. Michaelis, einem Professor für Philosophie an der damals neu gegründeten Universität Göttingen, angeregten, vom dänischen König Friedrich V. († 1766) finanzierten Expedition von sechs Personen der Jemen war, der am 27.12.1762 mit der Ankunft in al-Luḥayya erreicht wurde. Doch durch den Tod des Philologen und des Botanikers am 25.5. bzw. am 11.7.1763 entschloss man sich bereits am 24.8.1763, nach Indien abzusegeln, aber durch den Tod des Zeichners und des Assistenten nur wenige Tage später kamen nur noch Niebuhr und der Arzt am 11.9.1763 in Bombay in Indien an, und nachdem auch dieser im Februar 1764 verstarb, setzte Niebuhr als Einzelforscher die Reise durch den Vorderen Orient bis zu seiner Rückkehr im Herbst 1767 fort...
Rezension zu: Sara Stöcklin-Kaldewey, Kaiser Julians Gottesverehrung im Kontext der Spätantike

Von einem nachlassendem Interesse an der Gestalt des letzten Kaisers der konstantinischen Dynastie, der sich vom Christentum abwandte, kann wahrlich nicht gesprochen werden. Erst im vergangenen Jahr (2013) ist die Arbeit von Th. Nesselrath erschienen, die nach den Konzepten und Vorbildern der Repaganisierung des Reiches fragte. Nun hat S. Stöcklin-Kaldewey eine Dissertation vorgelegt, die von M. Wallraff (Universität Basel, Kirchen- und Theologiegeschichte; evang.) betreut wurde und die allein schon durch ihren Umfang (456 Seiten) beeindruckt. Die Biographie des Kaisers wird nur ganz sporadisch behandelt, seine politischen Maßnahmen werden ausgeblendet (vgl. S. 13). Umso mehr ist das Buch für Philosophen und Religionswissenschaftler eine anspruchsvolle, aber auch höchst bereichernde Lektüre. Die große Zahl an Werken, die aus der Feder des Kaisers erhalten ist, bietet zweifellos eine hervorragende Basis, die die Vf. im besten Sinne genutzt hat für ihre Frage nach Julians Gottesverehrung im Kontext der Spätantike. Wer das Buch gründlich liest, wird schnell feststellen, dass ihm hier mehr geboten wird, als der Titel vermuten lässt. Die Vf. sieht in Julian mit Recht „eine Schlüsselfigur für das Verständnis der religiösen Kultur der Spätantike“ (S.2) mit ihrer „wechselseitigen Durchdringung verschiedener religiöser Sphären“ (S.4)...

In data protection and the fight against cybercrime, European norms have been successfully globalized. The time is right to take the next step: Europe must now become the international normative leader for developing a new deal on internet governance. To ensure this, European powers should commit to rules that work in security, economic development and human rights on the internet and implement them in a reinvigorated IGF.

Dieses Working Paper zeigt Wege auf, wie völkerrechtlich verbindliche Regeln im Bereich der Cyber-Sicherheit entwickelt werden können. Wichtige Wegmarken können dabei nichtbindende Normen darstellen; auch aus Völkergewohnheitsrecht – besonders dem Kooperationsgebot – lassen sich präventive Schutzpflichten für Staaten (‚due diligence’) ableiten. Diesen präventiven Schutzpflichten müssen Staaten mit gemeinsamem Handeln zur Hebung von Cyber-Sicherheit gerecht werden. Um langfristig Rechts-sicherheit zu schaffen und Cyber-Sicherheit ganzheitlich zu fördern, führt aber kein Weg am Abschluss eines verbindlichen Übereinkommens über Cyber-Sicherheit vorbei.
Law is force of order. It reacts, usually with a necessary time delay, to technological pro-gress. Only twelve years after Samuel Morse presented the first workable telegraph sys-tem in New York in 1838 and six years after the first completed telegraph line from Wash-ington to Baltimore, central European states agreed on an international framework for tel-egraphs. It has been much more than twelve years since the technologies underlying the internet’s popularity today, such as the ‘World Wide Web’, were invented. No international framework has emerged, even though normative approaches abound. There are norms that are applied to the internet, but the recognition of the existence of an underlying, structuring order is missing. This motivates the present study.
Im Rahmen einer prospektiven Kohortenstudie wurde in der vorliegenden Arbeit die Epidemiologie und Ätiologie von Fieber während chemotherapiebedingter Neutropenie bei Patienten mit hämatologischen Neoplasien untersucht. Das diagnostische und therapeutische Vorgehen wurde beschrieben und die Übereinstimmung mit den Vorgaben einer hausinternen Leitlinie wurde evaluiert und der Verbrauch an antimikrobiellen Substanzen dokumentiert. Febrile Episoden konnten in 74% der Neutropenieepisoden dokumentiert werden; in 51% der Neutropenieepisoden kam es zu persistierendem Fieber ≥ 3 Tage. Eine dem Fieber zugrunde liegende Infektion wurde in 55% der Chemotherapiezyklen (Neutropeniedauer Median 16 Tage) mittels klinischer, apparativer und mikrobiologischer Diagnostik gefunden. Die häufigsten infektiösen Komplikationen waren Enteritiden mit Nachweis von C. difficile (11%), Enteritiden ohne Nachweis von C. difficile (29%), Pneumonien mit pilztypischen Infiltraten (12%) und Pneumonien mit unspezifischen Infiltraten (11%), sowie mikrobiologisch bestätigte Septikämien (18%). Im Vergleich zu Angaben in der Literatur wurde FUO in der vorliegenden Untersuchung häufiger diagnostiziert (11,8 Tage pro 1000 Patiententage vs. 8,2 Tage pro 1000 Patiententage). Die Gesamtmortalität des Kollektivs lag bei 7%. Die häufigste Todesursache war der septische Schock (8 von 5 Patienten; 63%), welcher sich auf dem Boden einer gramnegativen Septikämie mit multiresistenten Keimen entwickelt hatte (3 von 5 Septikämien; 60%). Die Mortalitätsrate war ähnlich hoch wie im Literaturvergleich bei Studien mit akuten myeloischen Leukämien. Prognoserelevante Faktoren, welche im Rahmen der vorliegenden Studie evaluiert wurden, sind durch schwer therapierbare Grunderkrankungen hervorgerufene prolongierte Neutropeniezeiten, sowie infektiöse Komplikationen, vor allem bei Nachweis multiresistenter Erreger.
Die Compliance bezüglich des diagnostischen und therapeutischen Vorgehens bei persistierendem Fieber betrug in der vorliegenden Arbeit zwischen 47% und 82%. Die Compliance mit den in der Leitlinie vorgeschlagenen antimikrobiellen prophylaktischen und initialen empirischen Therapien lag zwischen 83% und 85%. Der Gesamtverbrauch an antibiotischen und antimykotischen Substanzen war in der vorliegenden Studie höher als im Literaturvergleich (1647 DDD vs. 1140 DDD und 650 DDD vs. 430 DDD). Dabei fällt insbesondere der hohe Verbrauch an Reservemedikamenten gegen grampositive Bakterien auf. Allerdings fehlen aktuelle Vergleichsdaten vergleichbarer Patientenkollektive.
Bezüglich der Optimierung der diagnostischen Maßnahmen ist vor allem die Erhöhung der Sensitivität der Bronchoalveolären Lavage bei Pilzinfektionen, rechtzeitig und regelmäßig durchgeführte Surveillanceabstriche bei febriler Neutropenie, sowie die Diskussion der diagnostischen Ergebnisse und des weiteren therapeutischen Vorgehens mit infektiologischen Experten zu fordern. Patienten mit persistierendem Fieber - vor allem bei positivem Blutkulturnachweis - sollten, aufgrund des positiven prädiktiven Wertes erregerpositiver Blutkulturen für die Notwendigkeit einer intensivmedizinischen Betreuung und einer erhöhten Mortalität, Anlass sein, diagnostische und therapeutische Maßnahmen zu verbessern. Ziel ist es, durch verstärkten Einsatz diagnostischer Maßnahmen und Diskussion mikrobiologischer Befunde im Einzelfall, Reservesubstanzen seltener einzusetzen, gegebenenfalls Breitspektrum-Antibiotika oder ihre Kombinationen zu deeskalieren und somit die Kolonisation mit resistenten Erregern positiv zu beeinflussen, sowie das Ansprechen auf antimikrobielle Therapien zu optimieren.
Die Teilnahme an multizentrischen Surveillancestudien zur Inzidenzerfassung wichtiger, eindeutig definierter neutropener nosokomialer Infektionen wie radiologisch detektierter Pneumonien, mikrobiologisch bestätigter Septikämien und C. difficilepositiver Enteritiden, sowie zur Generierung von Antiinfektivaverbrauchszahlen, könnte helfen, die Beratung und Entscheidungsfindung am Krankenbett zu unterstützen. Auch könnten in diesem Rahmen Zahlen generiert werden, welche bisher zum Vergleich fehlten.
Die vorliegende Arbeit versteht sich im Rahmen der Erfassung von Ätiologie, Diagnostik und antimikrobieller Therapien bei febriler Neutropenie als Teil eines solchen Surveillanceprogrammes und hat somit eine Grundlagen für zukünftige Datenerhebungen geliefert.
Grafik-Hardware ist programmierbar geworden. Graphic Processing Units (GPUs) der neuen Generation wie der GeForce3 von NVIDIA enthalten Prozessoren, die es dem Software-Entwickler erlauben kurze Routinen auf der Grafik-Hardware auszuführen. Ich gebe in dieser Arbeit einen umfassenden Überblick über die Architekur und Leistungsfähigkeit dieser neuen Chipgeneration, zeige deren Stärken und Schwächen auf und diskutiere Verbesserungsvorschläge. Als Teil der Arbeit präsentiere ich einige von mir entwickelte Schattierungsverfahren, sowie eine Wassersimulation. Diese Demonstratoren sind darauf ausgerichtet vollständig auf den Prozessoren der neuen Grafikchip- Generation zu laufen. Als Antwort auf die Mängel der zur Zeit verfügbaren Application Programming Interfaces stelle ich ein alternatives Interface zur Steuerung der neuen GPUKomponenten vor, das insbesondere die Austauschbarkeit und Kombinierbarkeit von GPU-Programmen unterstützt.
Gegenstand der hier vorgestellten Arbeit ist eine Applikation für die virtuelle Realität (VR), die in der Lage ist, die Struktur eines beliebigen Textes als begehbare, interaktive Stadt zu visualisieren. Darüber hinaus bietet das Programm eine besondere Textsuche an, die so in anderen konventionellen Textverarbeitungsprogrammen nicht vorzufinden ist. Dank der strukturellen Analyse und der Verwendung einiger außergewöhnlicher Analysetools des TextImager [2], ermöglicht text2City nicht nur die Suche nach bestimmten Textmustern, sondern zum Beispiel auch die Bestimmung der Textebene (Wort, Satz, Absatz, etc.) und einiges mehr. Ein weiteres Feature ist die Kommunikationsverbindung zwischen dem TextAnnotator-Service [1] und text2City, die dem Benutzer die Möglichkeit zum Annotieren bietet, aber auch von anderen Personen durchgeführte Annotationen sofort sichtbar machen kann. Für die Ausführung des Programms ist eine der beiden VRBrillen, Oculus Rift oder HTC Vive, ein für VR geeigneter PC, sowie die Software Unity nötig.
Between 1996 and 2006 the vegetation succession in drift sands and in blown-out gravel-rich depressions, located in the nature reserve Hulshorsterzand in the central Netherlands, was studied. Within this Natura 2000 habitat (type 2330: inland dunes with open Corynephorus and Agrostis grasslands) so-called lichen steppes are included, famous for their biodiversity, both in flora (cryptogams), and in fauna. With multivariate analysis, the relation between the primary succession, the species composition of the vegetation and the soil quality was studied in three different biotopes i.e. 1. drift sand, 2. blownout gravel-rich depressions and 3. drift sand after management. With a total of up to 34 species, lichen diversity in the study area appeared to be high. Biotope 1, i.e. the pioneer vegetation with Corynephorus canescens on blowing sand, is rather scarce in lichens. Between 1996 and 2006 green algae, C. canescens and Polytrichum piliferum appeared, with the result that much of the former open drift sand area was covered by pioneer stages of the Spergulo-Corynephoretum. If sand stopped blowing in, a combination of P. piliferum with the neophyte Campylopus introflexus occurred. The terminal stages of lichen succession on drift sand, a vegetation with mainly Festuca ovina s.l., Deschampsia flexuosa and reindeer lichens (Cladonia subgen. Cladina), proved to be rather stable. Since 1996 the lichen steppes occurring in biotope 2 were increasingly grass-encroached. Where some sand was still blowing in, lichen diversity did not change much, but gradually C. introflexus increased. However, without sand blowingin, a decreasing lichen cover and loss of some Red List lichens occurred. In general, in this grass-rich vegetation Calluna vulgaris will germinate and gradually a dry heath might develop. However, where dynamics had stopped and Pinus sylvestris seedlings established, succession to a young forest started. Vegetation change from open sand to lichen-rich vegetation is clearly related to a decrease in pH, an increase in organic matter, in % total N and in % total P. The lichen composition is clearly related to this gradient. The applied restoration measures on steep sand dunes in the eastern area, both of cutting pine trees and removing topsoil down to the mineral soil layer, proved to be successful. The sand kept drifting and the pioneer community thriving, including the lichen Stereocaulon condensatum characteristic of pioneer conditions, while the neophytic moss hardly increased between 1996 and 2006.
With increasing distribution of endovascular stroke therapies, transient middle cerebral artery occlusion (tMCAO) in mice now more than ever depicts a relevant patient population with recanalized M1 occlusion. In this case, the desired therapeutic effect of blood flow restauration is accompanied by breakdown of the blood-brain barrier (BBB) and secondary reperfusion injury. The aim of this study was to elucidate short and intermediate-term transcriptional patterns and the involved pathways covering the different cellular players at the neurovascular unit after transient large vessel occlusion. To achieve this, male C57Bl/6J mice were treated according to an intensive post-stroke care protocol after 60 min occlusion of the middle cerebral artery or sham surgery to allow a high survival rate. After 24 h or 7 days, RNA from microvessel fragments from the ipsilateral and the contralateral hemispheres was isolated and used for mRNA sequencing. Bioinformatic analyses allowed us to depict gene expression changes at two timepoints of neurovascular post-stroke injury and regeneration. We validated our dataset by quantitative real time PCR of BBB-associated targets with well-characterized post-stroke dynamics. Hence, this study provides a well-controlled transcriptome dataset of a translationally relevant mouse model 24 h and 7 days after stroke which might help to discover future therapeutic targets in cerebral ischemia/reperfusion injury.

Dabei werden das Nachwirken von Imperialismus und Apartheid in Südafrika und Strategien der Dekolonialisierung in künstlerischen Praktiken thematisiert.

Das Ziel dieser Arbeit war die Evaluierung der Einsatzmöglichkeiten eines mikrostrukturierten Reaktorsystems in der heterogenen Katalyse. Hierzu wurde eine Reaktion herangezogen, welche typische Problemstellungen der heterogenen Katalyse abbildet. Zu diesen Problemen gehören Temperaturkontrolle, sichere Handhabung von explosiven Gasgemischen und das Erzielen von zufriedenstellenden Selektivitäten. Die Reaktion sollte außerdem bereits gut untersucht worden und die Prozessparameter aus der Literatur bekannt sein. Aus diesem Grund wurde die Partialoxidation von Ethen zu Ethenoxid an Silberkatalysatoren gewählt. Es konnte gezeigt werden, dass die Reaktion in einem Mikrostrukturreaktorsystem sicher durchführbar ist. Vor allem wurde an einer ganzen Reihe von Beispielen veranschaulicht, dass eine herausragende Eigenschaft des Mikrostrukturreaktors seine inhärente Explosionssicherheit ist. Gasgemische, welche sich mitten im explosiven Gemischbereich befanden, konnten bei Drücken von 2 bis 20 bar und Temperaturen von 230 bis 310 °C sicher gehandhabt werden. So konnte gezeigt werden, dass der Mikrostrukturreaktor sich dazu eignet Reaktionen mit explosiven Gasgemischen durchzuführen. Die Verwendung von Mikrostrukturreaktoren in der heterogenen Katalyse befindet sich noch im Anfangsstadium. Um Probleme bei der Übertragung von Katalysatorsystemen auf ein System mit Mikrostruktur zu vermeiden, erfolgte zunächst der Einsatz von Vollsilberkatalysatoren. Die Mikrostruktur wurde deshalb aus dem katalytisch aktiven Material selbst hergestellt. Die Herstellung wurde auf drei unterschiedliche Weisen (LIGA-, Ätz- und Sägeverfahren) durchgeführt. So konnte gezeigt werden, dass eine Kostenreduzierung bei der Darstellung von Mikrostrukturen möglich ist. Der Nachteil der Nutzung von Vollsilber war, dass sich deutlich schlechtere Selektivitäten bei der Partialoxidation von Ethen ergaben. Es konnte jedoch gezeigt werden, dass mit dem Mikrostrukturreaktor die Selektivitäten für Vollsilber im Schnitt 10 % über denen für Rohrreaktorexperimenten bei gleichen Umsätzen lagen. Die effektive Wärmeabführung und die homogene Verteilung der Wärme über den Mikrostrukturreaktor scheinen eine Verbesserung der Selektivität zu erbringen. Kinetische Untersuchungen zeigten, dass sowohl durch Anheben des Partialdrucks von Ethen als auch von Sauerstoff eine Erhöhung der Reaktionsgeschwindigkeit erzielt werden kann. Dabei wurde für Ethen eine formale Reaktionsordnung bei der Bildung von Ethenoxid von 0,53 gefunden, während sie für Sauerstoff 0,78 betrug. Mit diesen Untersuchungen wurde verdeutlicht, dass ein Erhöhen des Sauerstoffpartialdrucks einen positiven Einfluss auf die Selektivität hat. So konnte durch Anheben der Sauerstoffkonzentration von 5 %, wie es in industriellen Prozessen aus Sicherheitsgründen notwendig ist, auf bis zu 95 % eine Verbesserung der Selektivität von bis zu 15 % erzielt werden. Über diesen Sachverhalt wurde zwar bereits in der Literatur (16) berichtet, jedoch erfolgten die Untersuchungen hierfür unter Hochvakuumbedingungen. Der Mikrostrukturreaktor ermöglichte einen Nachweis dieses Phänomens auch unter Hochdruckbedingungen, wie sie für industrielle Reaktoren üblich sind. Damit konnte ein in der heterogenen Katalyse bekanntes Problem, nämlich die Übertragung von Erkenntnissen aus Ultrahochvakuumexperimenten auf Hochdruckbedingungen (pressure-gap), untersucht werden. Eine wissenschaftliche Prüfung, ob dem Ergebnis die gleichen Ursachen sowohl im Ultrahochvakuum als auch bei Hochdruckbedingungen zugrunde liegen, muss noch erfolgen. Es zeigte sich aber auch, dass durch eine Verweilzeiterhöhung keine weitere Verbesserung der Raum-Zeit-Ausbeute möglich ist. Vielmehr wurde klar, dass Reaktionsgeschwindigkeit und Selektivität mit längeren Verweilzeiten abnehmen. Als Grund hierfür konnte die Bildung von elementarem Kohlenstoff an der Silberoberfläche festgestellt werden. Aufgrund der Limitierung bei der Verweilzeit wurden maximale Umsätze von 24 % erzielt. Der Einsatz von 1,2-Dichlorethan als Oxidationsinhibitor für Vollsilber wurde ebenfalls untersucht. Dabei konnte die Selektivität auf bis zu 69 % gesteigert werden. Es erfolgte jedoch eine Einbuße an Aktivität von etwa 42 %. Es ist bekannt, dass die Oberflächenmorphologie von Silberkatalysatoren unter Reaktionsbedingungen starke Veränderungen erfährt. (68) Es wurde aufgezeigt, dass dies für die Oberfläche von mikrostrukturierten Silberfolien ebenfalls festzustellen ist. Dabei wurde gleichzeitig festgestellt, dass die Katalysatoren trotz unterschiedlicher Herstellungsmethoden und den daraus resultierenden unterschiedlichen Oberflächenmorphologien vergleichbare Aktivitäten aufweisen. Industriell verwendete Katalysatoren basieren auf alpha-Aluminiumoxid als Trägermaterial. Dabei wurde bereits seit vielen Jahren an Optimierungen des Katalysators gearbeitet. Durch das Einstellen der spezifischen Oberfläche und Partikelgröße des Silbers und den Einsatz von Alkali- und Erdalkalimetallen als Promotoren werden so Katalysatoren hergestellt, welche eine Selektivität von 80 % besitzen. Die Übertragung dieser Erkenntnisse auf ein Mikrostrukturreaktorsystem kann nicht ohne weiteres vorgenommen werden. Es wurden verschiedene Darstellungsmöglichkeiten für eine alpha-Aluminiumoxidschicht in einem Mikrostrukturreaktor untersucht. Dabei zeigte sich, dass nur die direkte Darstellung von alpha- Aluminiumoxid ohne Phasenumwandlung aus anderen Modifikationen erfolgversprechend ist. Eine Darstellung der Aluminiumoxidschicht durch Sol-Gel- oder CVD-Prozesse war nicht erfolgreich, da die für die Phasenumwandlung von gamma-Aluminiumoxid nach alpha-Aluminiumoxid notwendige Temperatur von 1100 °C die Ausbildung einer Eisenoxidschicht an der Oberfläche der mikrostrukturierten Edelstahlfolien zur Folge hatte. Diese eignete sich nicht als Träger. Alternativ wurde erfolgreich der Einsatz von aluminiumhaltigen Edelstählen untersucht. Diese bilden beim Ausheizen bei 1100 °C eine alpha-Aluminiumoxidschicht an der Oberfläche aus, welche mittels Sputtern mit Silber geträgert wurde. Katalytische Untersuchungen zeigten, dass mit dem Einsatz von alpha-Aluminiumoxidträgern eine Verbesserung der Selektivität im Vergleich zu Vollsilber von 17 % erreicht werden kann. Gleichzeitig konnte anhand eines Gegenüberstellens von katalytischen Daten mit TEM-Aufnahmen der Sputterschichten festgestellt werden, dass eine geschlossene Silberschicht an der Oberfläche notwendig ist, um eine zufriedenstellende Aktivität und Selektivität zu erzielen. Während bei Schichtdicken von 1 nm noch einzelne Silberinseln an der Oberfläche zu finden sind, liegt bei einer Schichtdicke von 5 nm eine fast geschlossene Silberschicht vor. Ein Anheben der Schichtdicke ergab keine weitere Verbesserung der Aktivität oder Selektivität. Dagegen ergab der Einsatz von 1,2-Dichlorethan eine weitere Steigerung der Selektivität auf 77 %. Industriell eingesetzte Rohrbündelreaktoren erreichen im Sauerstoffverfahren eine Selektivität von 80 %. Die hier erzielten 77 % Selektivität bei vergleichbaren Umsätzen zeigt, dass der Einsatz eines Mikrostrukturreaktors für die Synthese von Ethenoxid möglich ist, vor allem unter dem Gesichtspunkt, dass Potenzial für die Optimierung von Reaktoren und die Katalysatorpräparation besteht. Die Nutzung von Reaktionsbedingungen, wie Ethen in reinem Sauerstoff, und der daraus resultierenden Verbesserung für Aktivität und Selektivität, ermöglichen Raum-Zeit-Ausbeuten, die über denen von Industriereaktoren liegen. Ob Mikrostrukturreaktoren in industriellen Prozessen jemals eingesetzt werden, hängt allein von ökonomischen Faktoren ab. Dazu müsste die Selektivität über die bestehenden 80 % angehoben werden. Zur Zeit entfallen 80 % der Produktionskosten von Ethenoxid auf den Rohstoff Ethen, so dass jeder Prozentpunkt, um den die Selektivität angehoben werden könnte, eine deutliche Kosteneinsparung mit sich brächte und darüber entschiede, ob ein neuer Prozess eingeführt wird. Hierzu wäre es auch notwendig, die Kosten für die Produktion der Mikrostrukturreaktoren pro Volumeneinheit um mehrere Größenordnungen zu reduzieren. Außerdem müssten Lösungen entwickelt werden, welche die Peripherie des Reaktors betreffen, vor allem die Heizung und die Gasversorgung. Im Rahmen dieser Arbeit sollte überprüft werden, welche Leistungsfähigkeit ein Mikrostrukturreaktorprozess im Vergleich zu einem bestehenden Prozess besitzt. Es konnte dargestellt werden, dass Raum-Zeit-Ausbeuten über denen eines Industriereaktors erzielt werden können bei vergleichbareren Selektivitäten. Außerdem konnte gezeigt werden, dass der Mikrostrukturreaktor ein geeignetes Werkzeug ist, welches helfen kann, Reaktionen unter bisher nicht einfach zugänglichen Bedingungen durchzuführen.
Atovaquone is a substituted 2-hydroxynaphthoquinone that is used therapeutically to treat Plasmodium falciparum malaria, Pneumocystis carinii pneumonia, and Toxoplasma gondii toxoplasmosis. It is thought to act on these organisms by inhibiting the cytochrome bc1 complex. We have examined the interaction of atovaquone with the bc1 complex isolated from Saccharomyces cerevisiae, a surrogate, nonpathogenic fungus. Atovaquone inhibits the bc1 complex competitively with apparent Ki = 9 nm, raises the midpoint potential of the Rieske iron-sulfur protein from 285 to 385 mV, and shifts the g values in the EPR spectrum of the Rieske center. These results indicate that atovaquone binds to the ubiquinol oxidation pocket of the bc1 complex, where it interacts with the Rieske iron-sulfur protein. A computed energy-minimized structure for atovaquone liganded to the yeast bc1 complex suggests that a phenylalanine at position 275 of cytochrome b in the bovine bc1 complex, as opposed to leucine at the equivalent position in the yeast enzyme, is responsible for the decreased sensitivity of the bovine bc1 complex (Ki = 80 nm) to atovaquone. When a L275F mutation was introduced into the yeast cytochrome b, the sensitivity of the yeast enzyme to atovaquone decreased (Ki = 100 nm) with no loss in activity, confirming that the L275F exchange contributes to the differential sensitivity of these two species to atovaquone. These results provide the first molecular description of how atovaquone binds to the bc1 complex and explain the differential inhibition of the fungal versus mammalian enzymes.
A large reef manta ray (Manta alfredi) aggregation has been observed off the north Sudanese Red Sea coast since the 1950s. Sightings have been predominantly within the boundaries of a marine protected area (MPA), which was designated a UNESCO World Heritage Site in July 2016. Contrasting economic development trajectories have been proposed for the area (small-scale ecotourism and large-scale island development). To examine space-use, Wildlife Computers® SPOT 5 tags were secured to three manta rays. A two-state switching Bayesian state space model (BSSM), that allowed movement parameters to switch between resident and travelling, was fit to the recorded locations, and 50% and 95% kernel utilization distributions (KUD) home ranges calculated. A total of 682 BSSM locations were recorded between 30 October 2012 and 6 November 2013. Of these, 98.5% fell within the MPA boundaries; 99.5% for manta 1, 91.5% for manta 2, and 100% for manta 3. The BSSM identified that all three mantas were resident during 99% of transmissions, with 50% and 95% KUD home ranges falling mainly within the MPA boundaries. For all three mantas combined (88.4%), and all individuals (manta 1–92.4%, manta 2–64.9%, manta 3–91.9%), the majority of locations occurred within 15 km of the proposed large-scale island development. Results indicated that the MPA boundaries are spatially appropriate for manta rays in the region, however, a close association to the proposed large-scale development highlights the potential threat of disruption. Conversely, the focused nature of spatial use highlights the potential for reliable ecotourism opportunities.
Hintergrund: Einer der wichtigsten Gründe für das Therapieversagen von Zytostatika ist die MDR von malignen Zellen. Deren Modulation mit sogenannten Chemosen sitizern scheint ein erfolgversprechender Therapieansatz zu sein, so daß zahlreiche in vitro Studien initiiert wurden. Jedoch beachten diese Studien nur selten den Effekt der Proteinbindung auf die zelluläre Aufnahme von Zytostatika und der Wirkung von Chemosensitizern. Der Vorhersagewert der in vitro Tests für die in vivo Effektivität steht somit zur Diskussion. Die Ziele dieser Untersuchung waren es deshalb a) die Bindungscharakteristiken des Anthrazyklins IDA in HS und Zellkulturlösungen zu bestimmen, b) den Effekt der Proteinbindung, sowie des Chemosensitizers RVRP und des AkutPhaseProteins AGP, auf die Aufnahme und Retention von IDA in eine humane HL60 Leukämiezellinie zu untersuchen und c) die Bedeutung der Proteinbin dung für die in vivoAktivität von Zytostatika und Chemosensitizern zu analysieren. Methoden: Die Proteinbindung von IDA wurde mit Hilfe der Gleichgewichtsdialyse in HS und gebräuchlichen Zellkulturlösungen bestimmt. Die Zellaufnahme und Retention von IDA wurde in einer sensiblen und einer Pgpexprimierenden, resistenten humanen Leukämiezellinie (HL60 und HL60Vinc), sowie in Lymphozyten gesunder Probanden untersucht. Die Versuchsbedingungen wurden durch verschiedene Inkubationsmedien und die Zugabe von RVRP und AGP variiert. Die IDA Konzentration wurde spektrophotofluorometrisch analysiert. Ergebnisse: In den untersuchten Inkubationsmedien variierte die freie Fraktion von IDA um mehr als 7fach von ungefähr 60% in 15% FCS/PBS zu nur 8% in HS. In den Zellinien HL60 und HL60Vinc war die zelluläre Aufnahme von IDA in dem Medium mit niedrigen Proteinkonzentrationen ungefähr dreimal höher als in HS. Die Bedeutung von Pgp für das Resistenzverhalten der Leukämiezellinie zeigte sich durch die erniedrigte Akkumulation von IDA in den resistenten Zellen. RVRP konnte in allen Medien die Differenz der IDA Aufnahme und Retention zwischen der sensiblen und der resistenten Zellinie aufheben. Jedoch konnte RVRP nicht den Effekt von hohen Proteinkonzentrationen auf die IDA Aufnahme ausgleichen. In Anwesenheit von AGP konnte sowohl in den Medien mit hohen als auch niedrigen Protein konzentrationen nur eine sehr geringe Chemosensitizerwirkung nachgewiesen werden. Das Akkumulationsverhalten der Lymphozyten glich der sensiblen HL60 Zellinie. Schlußfolgerung: Proteinbindungsphänomene können die Aktivität von Zytostatika und Chemosensitizern erheblich beeinflussen. So ist in vitro die Serumproteinbindung eine wichtige Determinante für die zelluläre Aufnahme von IDA. Der Modulationseffekt von RVRP reagiert weniger sensitiv auf Unterschiede in der Gesamtproteinkonzentration, allerdings kann er durch erhöhte AGP Konzentrationen, wie sie bei Tumorpatienten vorkommen, erheblich beeinflußt werden. Bei der Interpretation von in vitro funktionellen Tests der zytostatischen Effektivität und Chemosensitizerwirkung sollte daher die Serumproteinbindung berücksichtigt und Untersuchungen in HS einbezogen werden.
Invasive fungal infections are associated with increased mortality in hematological patients. Despite considerable advances in antifungal therapy, the evaluation of suspected treatment failure is a common clinical challenge requiring extensive diagnostic testing to rule out potential causes, such as mixed infections. We present a 64-year-old patient with secondary AML, diabetes mellitus, febrile neutropenia, and sinusitis. While cultures from nasal tissue grew Aspergillus flavus, a microscopic examination of the tissue was suggestive of concomitant mucormycosis. However, fluorescence in situ hybridization (FISH) using specific probes targeting Aspergillus and Mucorales species ruled out mixed infection. This was confirmed by specific qPCR assays amplifying the DNA of Aspergillus, but not of Mucorales. These results provided a rational basis for step-down targeted therapy, i.e., the patient received posaconazole after seven days of calculated dual therapy with liposomal amphotericin B and posaconazole. Despite clinical response to the antifungal therapy, he died due to the progression of the underlying disease within two weeks after diagnosis of fungal infection. Molecular diagnostics applied to tissue blocks may reveal useful information on the etiology of invasive fungal infections, including challenging situations, such as with mixed infections. A thorough understanding of fungal etiology facilitates targeted therapy that may improve therapeutic success while limiting side effects.

Abstract
Objectives: Patients with open pulmonary tuberculosis (opTB) are subject to strict isolation rules. Sputum smear microscopy is used to determine infectivity, but sensitivity is lower than for culture. This study aimed to investigate the clinical relevance of this mismatch in contemporary settings.
Methods: Differential results between microscopy and culture were determined at the time of microscopic sputum conversion, from all patients with opTB between 01/2013 and 12/2017. In addition, data on HIV, multi/extensive drug-resistant TB status, time to smear- and cultural-negativity conversion were analyzed; and a Kaplan-Meier curve was developed.
Results: Of 118 patients with opTB, 58 had demographic data available for microbiological and clinical follow-up analysis; among these, 26 (44.8%) had still at least one positive culture result. Median time from opTB-treatment initiation to full microscopic sputum- or culture conversion, was 16.5 days (range 2-105), and 20 days (1-105), respectively (median difference: +3.5 days). Sixteen days after de-isolation, >90% had converted culturally. HIV- or multi/extensive drug-resistant TB status did not impact conversion time.
Conclusion: When patients with opTB were de-isolated after 3 negative sputum smear microscopy tests, a substantial part still revealed cultural growth of Mycobacterium tuberculosis complex, but it remains unclear, whether smear-negative and culturally-positive individuals on therapy are really infective. Thus, the clinical relevance of this finding warrants further investigation.
The taxonomic status of Alcyonium aurantiacum Quoy & Gaimard, 1833, an octocoral endemic to New Zealand, was reviewed through morpho-molecular data comparisons in an integrative approach. Molecular phylogenetic analyses (nuclear 28S and mitochondrial mtMutS) resolved New Zealand taxa as more closely related to other genera and nominal Alcyonium Linnaeus, 1758 from South America than to the genus’ North Atlantic type species. Due to low genetic variation, species delimitation relied predominantly on identifying consistent differences in sclerite and colony morphology. The former A. aurantiacum is reassigned to Kotatea gen. nov. as K. aurantiaca gen. et comb. nov. and seven new species are described in this genus (K. amicispongia gen. et sp. nov., K. lobata gen. et sp. nov., K. kapotaiora gen. et sp. nov., K. kurakootingotingo gen. et sp. nov., K. niwa gen. et sp. nov., K. raekura gen. et sp. nov., and K. teorowai gen. et sp. nov.). Three new species in Ushanaia gen. nov. are also described (U. ferruginea gen. et sp. nov., U. fervens gen. et sp. nov. and U. solida gen. et sp. nov. ). These descriptions increase our understanding of New Zealand’s endemic octocoral diversity and contribute to ongoing systematic revisions of Alcyonium.
Identifizierung und Charakterisierung von Liganden für Faktor VIII neutralisierende Antikörper

Das Fehlen von funktionellem Blutgerinnungsfaktor VIII (FVIII) in Hämophilie A- (HA-) Patienten wird durch Substitution mit FVIII-Präparaten therapiert. Die wesentlichste gegenwärtige Komplikation der FVIII-Ersatz-Therapie besteht in dem Auftreten von FVIII neutralisierenden Antikörpern (Inhibitoren) gegenüber exogenem FVIII. Diese können mittels verschiedener, kostenintensiver Therapien zur Induktion einer Immuntoleranz (ITI) mit unterschiedlichem Erfolg eliminiert werden. Für Patienten mit persistierenden Inhibitoren bedeuten diese nicht nur eine drastische Verminderung der Lebensqualität sondern ein lebensbedrohliches Szenario. Eine Liganden-vermittelte Blockierung von neutralisierenden anti-FVIII Antikörpern sowie die zielgerichtete Ansteuerung des Rezeptors FVIII-spezifischer Gedächtnis-B-Zellen stellen mögliche Ansätze zur Verwirklichung antigenspezifischer ITI-Strategien für eine dauerhafte, vollständige Eliminierung von FVIII-Inhibitoren dar. Zu diesem Zweck wurden in dieser Arbeit durch Screening von phagenpräsentierten, randomisierten Peptidbibliotheken mit Inhibitor-positiven Patientenplasmen Peptidliganden selektioniert. Diese wiesen eine spezifische Bindung von anti-FVIII Antikörpern in den verwendten Plasmen auf. Durch den Einsatz entsprechender Software konnten AS-Konsensusmotive der Peptidsequenzen möglichen, konformationellen, funktionellen Inhibitorepitopen in der A2- sowie C2-Domäne von FVIII zugeordnet werden. Die von in silico-Analysen vorgegebene Domänenspezifität der anti-FVIII Antikörper wurde in Bindungsstudien mit rekombinant exprimierten FVIII-Domänen verfiziert. Die korrespondierenden, synthetischen Peptidliganden blockierten die IgG-Bindung an FVIII und regenerierten partiell dessen Aktivität im Plasma. Die Peptide stellten funktionelle Mimotope der möglichen Inhibitorepitope in der A2- und C2-Domäne dar. Da FVIII neutralisierende Antikörper zumeist Epitope in beiden Domänen erkennen, wurden die Mimotope kombiniert, was in einer noch effektiveren Blockierung von FVIII-Inhibitoren resultierte. Weiterhin wiesen Mimotopkombinationen Kreuzreaktivität mit anti-FVIII IgG in heterologen Patientenplasmen auf. Durch Fusion der Peptide an die Multimerisierungsdomäne der alpha-Kette des humanen C4-Bindeproteins konnten in Zellkultur heptamere Proteine generiert werden. Gegenüber den synthetischen Peptiden wiesen die Multimere aufgrund ihrer Multivalenz sowie der strukturellen Integrität eine deutlich verbesserte Blockierung von anti-FVIII IgG auf. Das Multimerisierungskonzept erlaubte ferner die Kombination unterschiedlicher Peptidliganden in einem Heteromultimer, was anhand der selektierten, funktionellen Mimotope für mögliche A2- und C2-Epitope getestet wurde. Weiterhin zeichneten sich die Inhibitor-spezifischen Multimere gegenüber den synthetischen Peptiden durch deutlich verlängerte Halbwerstzeiten aus. In Präparationen peripherer mononuklearer Zellen (PBMCs) von Patienten färbten synthetische Peptide sowie Fluoreszenz-markierter FVIII B-Zellsubpopulationen mit einem Gedächntis-B-Zell Phänotyp (CD19+IgG+). Gedächtnis-B-Zellen in PBMCs wurden polyklonal stimuliert. Im ELISPOT-Verfahren konnten Tetanusspezifische, jedoch keine FVIII-spezifischen Zellen, detektiert werden. Im Gegensatz zu den verwandten Kontrollen bewirkte eine Präinkubation der Zellen mit dem Peptid 12C6, welches an das toxische D-AS-Peptid (KLAKLAK)2 gekoppelt war, allerdings eine Reduktion von anti-FVIII IgG in den Überständen stimulierter Zellen.
Sulphuric acid, ammonia, amines, and oxidised organics play a crucial role in nanoparticle formation in the atmosphere. In this study, we investigate the composition of nucleated nanoparticles formed from these compounds in the CLOUD chamber experiments at CERN. The investigation is carried out via analysis of the particle hygroscopicity, ethanol affinity, oxidation state, and ion composition. Hygroscopicity was studied by a hygroscopic tandem differential mobility analyser and a cloud condensation nuclei counter, ethanol affinity by an organic differential mobility analyser and particle oxidation level by a high-resolution time-of-flight aerosol mass spectrometer. The ion composition was studied by an atmospheric pressure interface time-of-flight mass spectrometer. The volume fraction of the organics in the particles during their growth from sizes of a few nanometers to tens of nanometers was derived from measured hygroscopicity assuming the Zdanovski-Stokes-Robinson relationship, and compared to values gained from the spectrometers. The ZSR-relationship was also applied to obtain the measured ethanol affinities during the particle growth, which were used to derive the volume fractions of sulphuric acid and the other inorganics (e.g. ammonium salts). In the presence of sulphuric acid and ammonia, particles with a mobility diameter of 150 nm were chemically neutralised to ammonium sulphate. In the presence of oxidation products of pinanediol, the organic volume fraction of freshly nucleated particles increased from 0.4 to ∼0.9, with an increase in diameter from 2 to 63 nm. Conversely, the sulphuric acid volume fraction decreased from 0.6 to 0.1 when the particle diameter increased from 2 to 50 nm. The results provide information on the composition of nucleated aerosol particles during their growth in the presence of various combinations of sulphuric acid, ammonia, dimethylamine and organic oxidation products.
Sulphuric acid, ammonia, amines, and oxidised organics play a crucial role in nanoparticle formation in the atmosphere. In this study, we investigate the composition of nucleated nanoparticles formed from these compounds in the CLOUD (Cosmics Leaving Outdoor Droplets) chamber experiments at CERN (Centre européen pour la recherche nucléaire). The investigation was carried out via analysis of the particle hygroscopicity, ethanol affinity, oxidation state, and ion composition. Hygroscopicity was studied by a hygroscopic tandem differential mobility analyser and a cloud condensation nuclei counter, ethanol affinity by an organic differential mobility analyser and particle oxidation level by a high-resolution time-of-flight aerosol mass spectrometer. The ion composition was studied by an atmospheric pressure interface time-of-flight mass spectrometer. The volume fraction of the organics in the particles during their growth from sizes of a few nanometers to tens of nanometers was derived from measured hygroscopicity assuming the Zdanovskii–Stokes–Robinson relationship, and compared to values gained from the spectrometers. Diese Abschlussarbeit zur dialektalen Gliederung des Zazaki besteht aus 2 Hauptteilen. Teil 1 beschreibt nach kurzer Darlegung und Auswertung der bisherigen Forschung die dialektale Struktur der Sprache und die Einteilung in Hauptdialekte. Teil 2 besteht aus Übersetzungen eines Beispieltextes in verschiedene Dialekte und Mundarten sowie aus Sprachkarten mit Isoglossen.
Hypoxic pulmonary vasoconstriction (HPV) redistributes pulmonary blood flow from areas of low oxygen partial pressure to areas of normal or relativity high oxygen availability, thus optimising the matching of perfusion to ventilation and preventing arterial hypoxemia. Generalised alveolar hypoxia results in a sustained increase in pulmonary artery pressure which in turn leads to structural changes in the walls of the pulmonary vasculature (pulmonary vascular remodelling). Recent findings have indicated a role for cytochrome P450 (CYP) epoxygenase-derived epoxyeicosatrienoic acids (EETs) in hypoxia-induced pulmonary vasoconstriction. Given that the intracellular concentration of EETs is determined by the soluble epoxide hydrolase (sEH), which metabolises EETs to their less active dihydroxyeicosatrienoic acids (DHETs), we assessed the influence of the sEH and EETs on pulmonary artery pressure, acute and chronic HPV, and pulmonary vascular remodelling in the mouse lung. In isolated lungs from wild-type mice, acute HPV was significantly increased by sEH inhibition, an effect abolished by pre-treatment with CYP epoxygenase inhibitors and the EET antagonist 14,15-EEZE. The acute hypoxia-induced vasoconstriction and EET production were greater in lungs from sEH-/- mice than from wild-type mice and sEH inhibition had no further effect on HPV in lungs from the former animals, while MSPPOH (CYP epoxygenase inhibitor) and 14,15-EEZE decreased the response. Exogenous application of 11,12-EET increased pulmonary artery pressure in a concentration-dependent manner and enhanced acute HPV in wild-type lungs, while 14,15-EET and 11,12-DHET were without significant effect on pulmonary artery pressure. 5-HT2A receptor antagonism or Rho kinase inhibition shifted the EET concentration-response curve to the right and abrogated the EET- and sEH inhibition-induced potentiation of acute hypoxic vasoconstriction. In lungs from wild-type and sEH-/- mice, hypoxic preconditioning (hypoxic ventilation for 10 minutes) enhanced the 5-HT response. 1-Adamantyl-3-cyclohexylurea (ACU), a sEH inhibitor, further amplified the hypoxia-induced 5-HT-hypersensitivity in wild-type mice. However, after hypoxic preconditioning, the sEH-/- lungs displayed a striking leftward shift in the 5-HT response. 11,12-EET can activate TRPC6 channels in endothelial cells by eliciting its translocation to the plasma membrane, more specifically to membrane domains enriched with the caveolae marker caveolin-1. This effect was also observed in rat pulmonary artery smooth muscle cells overexpressing the channel. Exposure of the latter cells to acute hypoxia also stimulated the intracellular translocation of TRPC6 to caveolae, an effect that was sensitive to the EET antagonist. The EET-induced translocation of TRPC6 channels was prevented by a 5-HT2A receptor antagonist but not by a Rho kinase inhibitor. Moreover, while acute hypoxia and 11,12-EET increased pulmonary pressure in lungs from TRPC6+/- mice, lungs from TRPC6-/- mice did not respond to either stimuli. These results indicate that the sEH and CYP-derived EETs are involved in acute HPV and that EET-induced pulmonary contraction under normoxic and hypoxic conditions involves a TRPC6 channel, a 5-HT2A receptor-dependent pathway and Rho kinase activation. In the second part of the study the role of the sEH in the development of pulmonary hypertension and vascular remodelling induced in mice by exposure to hypoxia (10% O2) for 21 days was analysed. In wild-type mice, chronic hypoxia decreased the pulmonary expression/activity of the sEH, induced right heart hypertrophy and erythropoiesis, and increased the number of partially and fully muscularised pulmonary resistance arteries (by 3-fold). Moreover, in HEK 293 cells, hypoxia (1% O2 up to 24 h) decreased sEH promoter activity by 50%. In isolated lungs, pre-exposure to chronic hypoxia significantly increased baseline perfusion pressures and potentiated the acute HPV. While an sEH inhibitor, ACU, potentiated acute HPV in lungs from mice maintained in normoxic conditions, it had no effect on HPV in lungs from mice exposed to hypoxia. The EET antagonist, 14,15-EEZE, abolished the sEH inhibitor-dependent increase in acute HPV in normoxic lungs and decreased HPV in chronic hypoxic lungs. Hypoxia-induced right heart hypertrophy and erythropoiesis were more pronounced in sEH-/- than in wild-type mice. Under normoxic and hypoxic conditions the muscularisation of resistance pulmonary arteries was greater in lungs from sEH-/- mice than in lungs from wild-type mice. sEH-/- mice also displayed an enhanced acute HPV, compared to that observed in wild-type mice and chronic exposure to hypoxia did not further potentiate acute HPV. However, in the presence of 14,15-EEZE responses returned to levels observed in normoxic lungs from wild-type animals. Furthermore, immunohistochemistry demonstrated an extensive expression of the sEH in the medial wall of pulmonary arteries from human donor lungs. Whereas sEH expression was not detectable in samples from pulmonary hypertension patients, indicating that the sEH is involved in hypoxia-induced pulmonary vascular remodelling and hypoxic pulmonary vasoconstriction. Taken together, the results presented in this thesis indicate that the expression/activity of the sEH is an important determinant of the magnitude of acute and chronic hypoxia-induced pulmonary vasoconstriction and pulmonary vascular remodelling by inactivating vasoconstrictor CYP-derived EETs. As sEH inhibitors are currently being developed for the treatment of human systemic hypertension, it should be noted that these compounds may even promote the development of pulmonary hypertension.
We present results for calculating fusion cross-sections using a new microscopic approach based on a time-dependent density-constrained DFT calculations. The theory is implemented by using densities and other information obtained from TDDFT time-evolution of the nuclear system as a constraint on the density for DFT calculations.
İsviçreli gazeteci, foto muhabiri, yazar, fotoğrafçı Annemarie Schwarzenbach, gazetecilik göreviyle Ekim 1933'te, Irak'ta sonlanacak doğu yolculuğuna çıkmıştır. Seyahatinin iki ayını Türkiye'de geçirmiştir. Gördüklerini, edindiği izlenimler çerçevesinde kaleme alarak, 1935 yılında "Winter in Vorderasien - Tagebuch einer Reise" başlığıyla yayınlatmıştır. Seyahat, Türkiye'de reform çalışmalarına ve Cumhuriyet'in onuncu yılına denk gelmektedir. Ancak aktarımlarına bakıldığında, eleştirel bakış açısı dikkat çekmektedir. Yürütülecek bu çalışma ile Schwarzenbach'ın Alman edebiyatına sunduğu Türkiye imgesinin anlaşılmasına katkı sağlanması amaçlanmıştır. Bu nedenle, seyahatname Türkiye'ye dair tasvirleri çerçevesinde ele alınacaktır.
Fırtına ve Coşku edebi akımı yazarlarından Heinrich Leopold Wagner'in "Kindermörderin" adlı eseri, akımın en önemli eserleri arasında gösterilmektedir. Wagner, eseri yayınlandığında, yazım stili ve konuları açıklıkla ele almış olması nedeniyle oldukça ağır eleştirilere maruz kalmıştır. Burjuva trajedisi türündeki eserinde, 18. yüzyıl toplum yaşamının en önemli konularından olan çocuk cinayeti motifini işlemiştir. Ancak, motifin karakteristik özelliği olan soylu bir erkekle saf burjuva kızı ilişkisini kurgularken, motifin ayrıntılarında değişikliğe gittiği görülmektedir. Bu bağlamda, kısaca Fırtına ve Coşku akımına ve 18. yüzyıl Alman toplumunda çocuk cinayeti olgusuna değinilecektir. Eser, dönem ve olguların arka planına dair verilen bu bilgilerden sonra, Fırtına ve Coşku akımının çocuk cinayeti motifini işleyişi ele alınacaktır. Ardından, Wagner'in, eserinde çocuk cinayeti motifini işlerken gittiği değişiklikler ve bu yolla ortaya koyduğu olgular saptanacaktır. Böylece, yürütülecek bu çalışmayla, çocuk cinayeti motifi konusunda yapılmış genel geçer tanımların dışına çıktığı ortaya konularak, Wagner'in eserine ve çocuk cinayeti motifi üzerine yapılacak araştırmalara katkı sağlanması amaçlanmıştır.
Epoxides and diols of polyunsaturated fatty acids (PUFAs) are bioactive and can influence processes such as tumor cell proliferation and angiogenesis. Studies with inhibitors of the soluble epoxide hydrolase (sEH) in animals overexpressing cytochrome P450 enzymes or following the systemic administration of specific epoxides revealed a markedly increased incidence of tumor metastases. To determine whether PUFA epoxides increased metastases in a model of spontaneous breast cancer, sEH-/- mice were crossed onto the polyoma middle T oncogene (PyMT) background. We found that the deletion of the sEH accelerated the growth of primary tumors and increased both the tumor macrophage count and angiogenesis. There were small differences in the epoxide/diol content of tumors, particularly in epoxyoctadecamonoenic acid versus dihydroxyoctadecenoic acid, and marked changes in the expression of proteins linked with cell proliferation and metabolism. However, there was no consequence of sEH inhibition on the formation of metastases in the lymph node or lung. Taken together, our results confirm previous reports of increased tumor growth in animals lacking sEH but fail to substantiate reports of enhanced lymph node or pulmonary metastases.
Der Name Engelbert Humperdinck wird von den meisten Musikinteressierten unwillkürlich mit seinem berühmtesten Werk "Hänsel und Gretel" oder allgemein mit dem Begriff "Märchenoper" assoziiert. Auch herausragende Musikwissenschaftler - wie z.B. Carl Dahlhaus im "Neuen Handbuch der Musikwissenschaft" - beschränken sich beim Stichwort "Humperdinck" in der Regel auf Erläuterungen zu "Hänsel und Gretel". Humperdinck hat in der Tat eine ganze Reihe von Märchenstoffen vertont: Er begann mit Liedern zu Märchenspielen, die seine Schwester Adelheid Wette für ihre Kinder geschrieben hatte ("Schneewittchen" 1888, "Hänsel und Gretel" 1890 und "Die sieben Geislein" 1895). Nach dem Erfolg der durchkomponierten Oper "Hänsel und Gretel" 1893 versuchte er sich an der Gattung Melodram ("Königskinder" 1895/97 nach einem Text von Elsa Bernstein), einer Mischform aus Singspiel und Melodram ("Dornröschen" 1902 nach einem Text von Elisabeth Filhés-Ebeling) und schließlich wieder an der durchkomponierten Märchenoper ("Königskinder" 1910). Außerdem schrieb er ein Vorspiel und zwei Musiknummern zu Maurice Maeterlincks "Der blaue Vogel" (1910-12).
Jedoch war dies nur ein Bereich der Musik für die Bühne, in dem er sich betätigte. Eine bedeutende Phase war für Humperdinck die Zusammenarbeit mit Max Reinhardt am Deutschen Theater und Alfred Halm am "Deutschen Schauspielhaus" in Berlin (von 1905 bis 1907), in der u.a. Bühnenmusik zu vier Shakespeareschen Werken entstand ("Der Kaufmann von Venedig", "Der Sturm", "Das Wintermärchen" und "Was ihr wollt"). In seiner letzten Schaffensperiode konnte Humperdinck sich verstärkt der Gattung widmen, die ihm schon vor Hänsel und Gretel als Ausweg aus der Wagner-Nachfolge-Lethargie erschienen war, der komischen Oper ("Heirat wider Willen" 1905, "Die Marketenderin" 1914, "Gaudeamus" 1919).
In diesem Beitrag soll "Dornröschen" genauer betrachtet werden - ein Märchenspiel, das im Unterschied zu den beiden Märchenopern Humperdincks wenig Erfolg hatte und in der Literatur bisher so gut wie gar nicht behandelt wurde. Wie ist das Werk entstanden, was ist das Besondere an ihm, und welche Gründe gibt es für sein Scheitern auf der Bühne? Zunächst soll je doch ein kurzer Überblick über Inhalt, Besetzung und Form in das Werk ein führen...
„Zu viel Musik“ lautet der Titel eines Aufsatzes des Pianisten, Komponisten, Dirigenten und Musikschriftstellers Ferdinand Hiller (1811-1885) über die Musikbelästigung eines musikalischen Bürgers durch unterschiedlichste Quellen in seinem Umfeld im Laufe eines Tages, sicherlich auf eigenen Erfahrungen fußend.1 Er wird als Titel für diesen Beitrag im übertragenen Sinne zitiert, weil der Nachlass Hiller in der Universitätsbibliothek Frankfurt am Main beim ersten Sichten bisher sehr unübersichtlich erschien. Der Nachlass umfasst 74 Bände Musikhandschriften, davon 21 Sammelhandschriften, dazu 9 Mappen mit Stimmen, 59 Bände Musikdrucke eigener Werke, davon 29 Sammelbände, dazu eine dicke Mappe mit Stimmen. Weitere Handschriften wurden nachträglich erworben. In verschiedenen Sammlungen sind darüber hinaus Briefe und Bildnisse Hillers überliefert.
Dieser Beitrag hat das Ziel, eine strukturierte Übersicht über die enthaltenen Werke zu geben: Der Inhalt der im Druck überlieferten Sammelbände wird anhand einer Tabelle dargestellt. Den Hauptteil dieses Beitrages bildet eine nach musikalischen Gattungen gegliederte Liste der in Frankfurt überlieferten Musikautographe, die als Register zu dem mittler weile digital verfügbaren Nachlassverzeichnis von Christine Ihl genutzt werden kann.
Zwei große Theaterzettelsammlungen mit über 100.000 Stück sind seit dem ersten Drittel des 20. Jahrhunderts in der damaligen Stadt- und heutigen Universitätsbibliothek Frankfurt am Main beheimatet und machen sie in dieser Hinsicht zu einer bedeutenden theaterhistorischen Institution im deutschsprachigen Raum: die Sammlung der Städtischen Bühnen Frankfurt am Main und ihrer Vorläufer sowie die Sammlung des Manskopfschen Musik- und Theaterhistorischen Museums...
Exzellenz ist in der Frankfurter Universität nicht nur in den Bereichen Naturwissenschaften, Gesellschaftswissenschaften und Wirtschaftwissenschaften zu finden. Eine Reihe historischer Sammlungen gibt ihr eine einzigartige Prägung und hebt sie von anderen Universitäten ab. Zu diesen gehört die Musik- und Theatersammlung der Universitätsbibliothek, die viertgrößte Musik- und größte Theatersammlung in deutschen wissenschaftlichen Bibliotheken (www.ub.uni-frankfurt.de/musik.html).
