Genetic data in studies of systematics of Amazonian amphibians frequently reveal that purportedly widespread single species in reality comprise species complexes. This means that real species richness may be significantly higher than current estimates. Here we combine genetic, morphological, and bioacoustic data to assess the phylogenetic relationships and species boundaries of two Amazonian species of the Dendropsophus leucophyllatus species group: D. leucophyllatus and D. triangulum. Our results uncovered the existence of five confirmed and four unconfirmed candidate species. Among the confirmed candidate species, three have available names: Dendropsophus leucophyllatus, Dendropsophus triangulum, and Dendropsophus reticulatus, this last being removed from the synonymy of D. triangulum. A neotype of D. leucophyllatus is designated. We describe the remaining two confirmed candidate species, one from Bolivia and another from Peru. All confirmed candidate species are morphologically distinct and have much smaller geographic ranges than those previously reported for D. leucophyllatus and D. triangulum sensu lato. Dendropsophus leucophyllatus sensu stricto occurs in the Guianan region. Dendropsophus reticulatus comb. nov. corresponds to populations in the Amazon basin of Brazil, Ecuador, and Peru previously referred to as D. triangulum. Dendropsophus triangulum sensu stricto is the most widely distributed species; it occurs in Amazonian Ecuador, Peru and Brazil, reaching the state of Pará. We provide accounts for all described species including an assessment of their conservation status.
Herb induced liver injury (HILI) and drug induced liver injury (DILI) share the common characteristic of chemical compounds as their causative agents, which were either produced by the plant or synthetic processes. Both, natural and synthetic chemicals are foreign products to the body and need metabolic degradation to be eliminated. During this process, hepatotoxic metabolites may be generated causing liver injury in susceptible patients. There is uncertainty, whether risk factors such as high lipophilicity or high daily and cumulative doses play a pathogenetic role for HILI, as these are under discussion for DILI. It is also often unclear, whether a HILI case has an idiosyncratic or an intrinsic background. Treatment with herbs of Western medicine or traditional Chinese medicine (TCM) rarely causes elevated liver tests (LT). However, HILI can develop to acute liver failure requiring liver transplantation in single cases. HILI is a diagnosis of exclusion, because clinical features of HILI are not specific as they are also found in many other liver diseases unrelated to herbal use. In strikingly increased liver tests signifying severe liver injury, herbal use has to be stopped. To establish HILI as the cause of liver damage, RUCAM (Roussel Uclaf Causality Assessment Method) is a useful tool. Diagnostic problems may emerge when alternative causes were not carefully excluded and the correct therapy is withheld. Future strategies should focus on RUCAM based causality assessment in suspected HILI cases and more regulatory efforts to provide all herbal medicines and herbal dietary supplements used as medicine with strict regulatory surveillance, considering them as herbal drugs and ascertaining an appropriate risk benefit balance.
Natural killer (NK) cells play an important role following allogeneic hematopoietic stem cell transplantation (HSCT) exerting graft-versus-leukemia/tumor effect and mediating pathogen-specific immunity. Although NK cells are the first donor-derived lymphocytes reconstituting post-HSCT, their distribution of CD56++CD16− (CD56bright), CD56++CD16+ (CD56intermediate=int), and CD56+CD16++ (CD56dim) NK cells is explicitly divergent from healthy adults, but to some extent comparable to the NK cell development in early childhood. The proportion of CD56bright/CD56int/CD56dim changed from 15/8/78% in early childhood to 6/4/90% in adults, respectively. Within this study, we first compared the NK cell reconstitution post-HSCT to reference values of NK cell subpopulations of healthy children. Afterward, we investigated the reconstitution of NK cell subpopulations post-HSCT in correlation to acute graft versus host disease (aGvHD) and chronic graft versus host disease (cGvHD) as well as to viral infections. Interestingly, after a HSCT follow-up phase of 12 months, the distribution of NK cell subpopulations largely matched the 50th percentile of the reference range for healthy individuals. Patients suffering from aGvHD and cGvHD showed a delayed reconstitution of NK cells. Remarkably, within the first 2 months post-HSCT, patients suffering from aGvHD had significantly lower levels of CD56bright NK cells compared to patients without viral infection or without graft versus host disease (GvHD). Therefore, the amount of CD56bright NK cells might serve as an early prognostic factor for GvHD development. Furthermore, a prolonged and elevated peak in CD56int NK cells seemed to be characteristic for the chronification of GvHD. In context of viral infection, a slightly lower CD56 and CD16 receptor expression followed by a considerable reduction in the absolute CD56dim NK cell numbers combined with reoccurrence of CD56int NK cells was observed. Our results suggest that a precise analysis of the reconstitution of NK cell subpopulations post-HSCT might indicate the occurrence of undesired events post-HSCT such as severe aGvHD.values
BACKGROUND: Time-limited, early-life exposures to institutional deprivation are associated with disorders in childhood, but it is unknown whether effects persist into adulthood. We used data from the English and Romanian Adoptees study to assess whether deprivation-associated adverse neurodevelopmental and mental health outcomes persist into young adulthood.
METHODS: The English and Romanian Adoptees study is a longitudinal, natural experiment investigation into the long-term outcomes of individuals who spent from soon after birth to up to 43 months in severe deprivation in Romanian institutions before being adopted into the UK. We used developmentally appropriate standard questionnaires, interviews completed by parents and adoptees, and direct measures of IQ to measure symptoms of autism spectrum disorder, inattention and overactivity, disinhibited social engagement, conduct or emotional problems, and cognitive impairment (IQ score <80) during childhood (ages 6, 11, and 15 years) and in young adulthood (22-25 years). For analysis, Romanian adoptees were split into those who spent less than 6 months in an institution and those who spent more than 6 months in an institution. We used a comparison group of UK adoptees who did not experience deprivation. We used mixed-effects regression models for ordered-categorical outcome variables to compare symptom levels and trends between groups.
FINDINGS: Romanian adoptees who experienced less than 6 months in an institution (n=67 at ages 6 years; n=50 at young adulthood) and UK controls (n=52 at age 6 years; n=39 at young adulthood) had similarly low levels of symptoms across most ages and outcomes. By contrast, Romanian adoptees exposed to more than 6 months in an institution (n=98 at ages 6 years; n=72 at young adulthood) had persistently higher rates than UK controls of symptoms of autism spectrum disorder, disinhibited social engagement, and inattention and overactivity through to young adulthood (pooled p<0·0001 for all). Cognitive impairment in the group who spent more than 6 months in an institution remitted from markedly higher rates at ages 6 years (p=0·0001) and 11 years (p=0·0016) compared with UK controls, to normal rates at young adulthood (p=0·76). By contrast, self-rated emotional symptoms showed a late-onset pattern with minimal differences versus UK controls at ages 11 years (p=0·0449) and 15 years (p=0·17), and then marked increases by young adulthood (p=0·0005), with similar effects seen for parent ratings. The high deprivation group also had a higher proportion of people with low educational achievement (p=0·0195), unemployment (p=0·0124), and mental health service use (p=0·0120, p=0·0032, and p=0·0003 for use when aged <11 years, 11-14 years, and 15-23 years, respectively) than the UK control group. A fifth (n=15) of individuals who spent more than 6 months in an institution were problem-free at all assessments.
INTERPRETATION: Notwithstanding the resilience shown by some adoptees and the adult remission of cognitive impairment, extended early deprivation was associated with long-term deleterious effects on wellbeing that seem insusceptible to years of nurturance and support in adoptive families.

Peripheral tolerance is an important mechanism by which the immune system can guarantee a second line of defense against autoreactive T and B cells. One autoimmune disease that is related to a break of peripheral tolerance is diabetes mellitus type 1. Using the RIP-GP mouse model, we analyzed the role of the spleen and lymph nodes (LNs) in priming CD8+ T cells and breaking peripheral tolerance. We found that diabetes developed in splenectomized mice infected with the lymphocytic choriomeningitis virus (LCMV), a finding showing that the spleen was not necessary in generating autoimmunity. By contrast, the absence of LNs prevented the priming of LCMV-specific CD8+ T cells, and diabetes did not develop in these mice. Additionally, we found that dendritic cells are responsible for the distribution of virus in secondary lymphoid organs, when LCMV was administered intravenously. Preventing this distribution with the sphingosine-1-phosphate receptor antagonist FTY720 inhibits the transport of antigen to peripheral LNs and consequently prevented the onset of diabetes. However, in case of subcutaneous infection, administration of FTY720 could not inhibit the onset of diabetes because the viral antigen is already presented in the peripheral LNs. These findings demonstrate the importance of preventing the presence of antigen in LNs for maintaining tolerance.
Background. Colorectal cancers (CRC) shed DNA into blood circulation. There is growing evidence that the analysis of circulating tumor DNA can be effectively used for monitoring of disease, to track tumor heterogeneity and to evaluate response to treatment. Case Presentation. Here, we describe two cases of patients with advanced CRC. The first case is about a patient with no available tissue for analysis of RAS mutation status. Liquid biopsy revealed RAS-wild-type and the therapy with anti-EGFR (epidermal growth factor receptor) monoclonal antibody cetuximab could be initiated. In the second case, the mutational profile of a patient with initial wild-type RAS-status was continually tracked during the course of treatment. An acquired KRAS exon 3 mutation was detected. The number of KRAS mutated fragments decreased continuously after the discontinuation of the therapy with EGFR-specific antibodies. Conclusion. Liquid biopsy provides a rapid genotype result, which accurately reproduces the current mutation status of tumor tissue. Furthermore, liquid biopsy enables close monitoring of the onset of secondary resistance to anti-EGFR therapy.
This introductory analysis on the subject of werewolves in the Greek and Roman worlds in its legendary, mythical, scientific and medical dimension emphasizes an intrinsic combination of negative and positive aspects, human and non-human factors, and ancient and modern components, laying the groundwork for the study of the gendered duplicity of the werewolf's Self in the modern and contemporary literature of southern and northern Italy. In this presentation of the werewolf motif on the Italian literary panorama from the 19th to the 21st century through an overview of short stories and novels, we will examine the writers who have combined ancient rural legends with metropolitan reveries to underscore the complexity and obscure double life of the werewolf.
Bei der Schlussszene in 'Nathan der Weise', in der auf Lessings Regieanweisung hin "Unter stummer Wiederholung allseitiger Umarmungen" der Vorhang fällt, wird das Toleranzpostulat der Ringparabel als wirksam besiegelt, obwohl die Erwartung an eine konventionelle Auflösung im Liebesglück enttäuscht wird. Die allseitige Umarmung erscheint manchem_r irritierend oder gar befremdend. Es stellt sich leicht ein Unbehagen ein. Doch unterliegt dem so tolerant gelösten Religionsstreit ein ethnographisches Tableau, auf dessen Basis die Differenz durch Transkulturalität bewältigt wird. Lessing klärt die in der Aufklärung aufkommende ethnografische Konstruktion von Differenz auf, indem er die religiöse und kulturelle Grenzziehung durch die Erzählung zweier Vorgeschichten ihrer essentiellen Grundlage beraubt. Zwar enden diese beide tragisch, denn Nathans Familie wird von antisemitischen Kreuzzugshorden ermordet und die Eltern des Geschwisterpaares sterben im Kreuzzug, doch die Kinder der transkulturellen Familiengründung, Recha/Blanda und der Tempelherr, überleben. Recha, die als Pfand der Tugend von Hand zu Hand geht, wird nicht nur aus den Flammen, sondern auch aus dem Zwiespalt der Loyalitäten gerettet. In Recha wird Transkulturalität zur Gestalt. Nicht nur auf ihrem Körper sondern auch in ihrem Geist oszilliert die Kommunikation und wird die Blockade zwischen den Kulturen überwunden. Dabei bleibt sie jedoch als machtlose weibliche Figur das zirkulierende Objekt der Begierde. Der Tempelherr - wie Recha ein Waise - durchläuft einen Bildungsprozess, in dem er seine Position ständig neu finden muss und Fragen der Transkulturalität ausgehandelt werden. Das happy-end, in dem sich Recha und der Tempelherr als Geschwister und verwaiste Kinder einer Deutschritterin und eines Bruders des Sultans in den Armen Nathans, des Sultans und dessen Schwester wiederfinden, entspricht weder dem romantischem Muster noch der Erwartung eines tragischen Verlusts. Nein, 'Nathan der Weise' ist nicht als Trauerspiel, auch nicht als Lustspiel angelegt. Das dramatische Gedicht in fünf Aufzügen setzt dagegen zwei Erzählungen ins Verhältnis - die über Toleranz und die über Transkulturalität. Während Toleranz als Gebot in der Ringparabel ausgegeben wird, wird Transkulturalität in Lessings multikulturellem Jerusalem des 11. Jahrhunderts, wenn auch unter klaren Machtverhältnissen, gelebt. Glücklich ausgehen kann das Drama nur, weil Lessing so der Transkulturalität durch Blutsbande eine tragfähige Basis schafft. Meine These ist, dass Lessing einen solchen Schluss schreiben musste, wenn er das in der Ringparabel entwickelte Gebot der Toleranz als reale Chance verwirklicht sehen wollte. Aus postkolonialer Sicht stellt sich für die heutige Rezeption die Frage, wie notwendig das Konzept der Kreolisierung für die Entwicklung einer toleranten Gesellschaft ist.
Nolan brings a fairly recognizable style to all his movies, often described as "dark" or "gritty." The tone is relentlessly serious, and the narratives are infused with the ambiguity and pessimism of film noir. His protagonists are, without exception, tortured, obsessed men, struggling with the loss of loved ones or past mistakes. Not only do these men face an uncaring world with murky morality, their sense of self is also unstable. To go along with the faulty memories and self-deceptions of his characters, Nolan also has a knack for misleading his audience with convoluted narratives. As Fisher puts it, he specializes in puzzles that can't be solved. 'Inception', to my mind, makes an excellent showcase for all these themes. Therefore, through an examination of its use of space and architecture as metaphors for the mind, I aim to determine the concept of the malleable self that underlies all of Nolan's movies.
Der vorliegende Artikel beschäftigt sich mit dem im Westen noch weitgehend unbekannten Genre des chinesischen Kriminalromans "gongàn xiaoshuo", sowie dessen produktiver Rezeption durch den niederländischen Diplomaten und Schriftsteller Robert H. van Gulik (*09.08.1910, †24.09.1967). Als Verständnisgrundlage sollen dabei zunächst einige grundsätzliche Ausführungen und Erläuterungen zu besagtem Genre getätigt werden. Danach wird sich der Text van Guliks "Richter Di" zuwenden, jener Romanreihe, der der Sinologe seine bis heute anhaltende Bekanntheit, auch über wissenschaftliche Kreise hinaus, verdankt. Um den Rahmen dieser Aufsatzes nicht zu sprengen, soll sich jedoch vorwiegend auf die durch van Gulik angefertigte und veröffentlichte Übersetzung des ursprünglichen chinesischen Originals, 'Wǔ Zétian sì dà qí àn' ( 武則天四大奇案 , "Vier außerordentlich seltsame Fälle in der Regierungszeit der Kaiserin Wu") (auch: 'Dee Goong An' ( 狄公案, "Richter Dis Fälle")), sowie den dritten Roman der Reihe, 'Geisterspuk in Peng-lai', konzentriert werden, da in diesem der Beginn der Karriere Richter Dis beschrieben wird. Auf diese Weise kann sowohl die besondere Form des chinesischen Kriminalromans - die van Gulik in seinen übrigen "Richter Di"- Romanen übernimmt -, wie auch das tradierte China-Bild und die Anklänge konfuzianischer Philosophie in angemessenem Umfang bearbeitet werden. Die in den Romanen dargestellten Formen des Alltagslebens, die Regierungsstrukturen, die politischen Verschwörungen und die Angriffe von außen, sind historisch korrekt beschrieben und gewähren somit einen faszinierenden Einblick in die Welt des historischen Chinas.
Die Bobrowski-Bibliographie versucht, möglichst vollständig und aktuell sämtliche Publikationen von und über Johannes Bobrowski bzw. über sein Werk zu erfassen. Um die Bibliographie überschaubar zu halten, wurden allerdings Einzelveröffentlichungen von Gedichten, Prosatexten, Rezensionen, Vor- und Nachworten, Interviews und Selbstzeugnissen von Bobrowski in 'deutscher Sprache' nicht aufgenommen. Diese Texte sind in den Gesammelten Werken leicht zugänglich und der Ort ihrer jeweiligen Erstveröffentlichung kann den beiden Erläuterungsbänden von Eberhard Haufe und Holger Gehle (= Gesammelte Werke Bnd. V und VI) entnommen werden.

Die vorliegende Ergänzung baut auf der 'Johannes-Bobrowski-Bibliographie von 2002' auf. Sie folgt dieser in Herangehensweise und Gliederung. Neben einigen dort nicht aufgeführten älteren Titeln enthält sie vor allem solche wissenschaftlichen Arbeiten zu und über Johannes Bobrowski, die zwischen 2002 und 2016 publiziert worden sind. Nicht systematisch erfasst wurden Neuausgaben und Übersetzungen seines Werkes sowie Miszellen, Zeitungsartikel und künstlerische Resonanzen.

We study platform design in online markets in which buying involves a (non-monetary) cost for consumers caused by privacy and security concerns. Firms decide whether to require registration at their website before consumers learn relevant product information. We derive conditions under which a monopoly seller benefits from ex ante registration requirements and demonstrate that the profitability of registration requirements is increased when taking into account the prospect of future purchases or an informational value of consumer registration to the
rm. Moreover, we consider the effectiveness of discounts (store credit) as a means to influence the consumers-registration decision. Finally, we con
rm the profitability of ex ante registration requirements in the presence of price competition.

Mean age of stratospheric air can be derived from observations of sufficiently long lived trace gases with approximately linear trends in the troposphere. Mean age can serve as a tracer to investigate stratospheric transport and long term changes in the strength of the overturning Brewer-Dobson circulation of the stratosphere. For this purpose, a low-cost method is required in order to allow for regular observations up to altitudes of about 30 km. Despite the desired low costs, high precision and accuracy are required in order to allow determination of mean age. We present balloon borne AirCore observations from two mid latitude sites: Timmins in Ontario/Canada and Lindenberg in Germany. During the Timmins campaign five AirCores sampled air in parallel from a large stratospheric balloon and were analysed for CO2, CH4 and partly CO. We show that there is good agreement between the different AirCores (better than 0.1 %) especially when vertical gradients are small. The measurements from Lindenberg were performed using small low-cost balloons and yielded very comparable results. We have used the observations to extend our long term data set of mean age observations at Northern Hemi-sphere mid latitudes. The time series now covers more than 40 years and shows a small, statis-tically not significant positive trend of 0.15 ± 0.18 years/decade. This trend is slightly smaller than the previous estimate of 0.24 ± 0.22 years/decade which was based on observations up to the year 2006. These observations are still in contrast to strong negative trends of mean age as derived from some model calculations.
Possible effects of clothianidin seed-treated oilseed rape on honey bee colonies were investigated in a large-scale monitoring project in Northern Germany, where oilseed rape usually comprises 25–33 % of the arable land. For both reference and test sites, six study locations were selected and eight honey bee hives were placed at each location. At each site, three locations were directly adjacent to oilseed rape fields and three locations were situated 400 m away from the nearest oilseed rape field. Thus, 96 hives were exposed to fully flowering oilseed rape crops. Colony sizes and weights, the amount of honey harvested, and infection with parasites and diseases were monitored between April and September 2014. The percentage of oilseed rape pollen was determined in pollen and honey samples. After oilseed rape flowering, the hives were transferred to an extensive isolated area for post-exposure monitoring. Total numbers of adult bees and brood cells showed seasonal fluctuations, and there were no significant differences between the sites. The honey, which was extracted at the end of the exposure phase, contained 62.0–83.5 % oilseed rape pollen. Varroa destructor infestation was low during most of the course of the study but increased at the end of the study due to flumethrin resistance in the mite populations. In summary, honey bee colonies foraging in clothianidin seed-treated oilseed rape did not show any detrimental symptoms as compared to colonies foraging in clothianidin-free oilseed rape. Development of colony strength, brood success as well as honey yield and pathogen infection were not significantly affected by clothianidin seed-treatment during this study.
This study was part of a large-scale monitoring project to assess the possible effects of Elado® (10 g clothianidin & 2 g β-cyfluthrin/kg seed)-dressed oilseed rape seeds on different pollinators in Northern Germany. Firstly, residues of clothianidin and its active metabolites thiazolylnitroguanidine and thiazolylmethylurea were measured in nectar and pollen from Elado®-dressed (test site, T) and undressed (reference site, R) oilseed rape collected by honey bees confined within tunnel tents. Clothianidin and its metabolites could not be detected or quantified in samples from R fields. Clothianidin concentrations in samples from T fields were 1.3 ± 0.9 μg/kg and 1.7 ± 0.9 μg/kg in nectar and pollen, respectively. Secondly, pollen and nectar for residue analyses were sampled from free flying honey bees, bumble bees and mason bees, placed at six study locations each in the R and T sites at the start of oilseed rape flowering. Honey samples were analysed from all honey bee colonies at the end of oilseed rape flowering. Neither clothianidin nor its metabolites were detectable or quantifiable in R site samples. Clothianidin concentrations in samples from the T site were below the limit of quantification (LOQ, 1.0 µg/kg) in most pollen and nectar samples collected by bees and 1.4 ± 0.5 µg/kg in honey taken from honey bee colonies. In summary, the study provides reliable semi-field and field data of clothianidin residues in nectar and pollen collected by different bee species in oilseed rape fields under common agricultural conditions.
The most frequently used parameters to describe the barrier properties of endothelial cells (ECs) in vitro are (i) the macromolecular permeability, indicating the flux of a macromolecular tracer across the endothelium, and (ii) electrical impedance of ECs grown on gold-film electrodes reporting on the cell layer's tightness for ion flow. Due to the experimental differences between these approaches, inconsistent observations have been described. Here, we present the first direct comparison of these assays applied to one single cell type (human microvascular ECs) under the same experimental conditions. The impact of different pharmacological tools (histamine, forskolin, Y-27632, blebbistatin, TRAP) on endothelial barrier function was analyzed by Transwell(®) tracer assays and two commercial impedance devices (xCELLigence(®), ECIS(®)). The two impedance techniques provided very similar results for all compounds, whereas macromolecular permeability readings were found to be partly inconsistent with impedance. Possible reasons for these discrepancies are discussed. We conclude that the complementary combination of both approaches is highly recommended to overcome the restrictions of each assay. Since the nature of the growth support may contribute to the observed differences, structure-function relationships should be based on cells that are consistently grown on either permeable or impermeable growth supports in all experiments.
We study exchangeable coalescent trees and the evolving genealogical trees in models for neutral haploid populations.
We show that every exchangeable infinite coalescent tree can be obtained as the genealogical tree of iid samples from a random marked metric measure space when the marks are added to the metric distances. We apply this representation to generalize the tree-valued Fleming-Viot process to include the case with dust in which the genealogical trees have isolated leaves.
Using the Donnelly-Kurtz lookdown approach, we describe all individuals ever alive in the population model by a random complete and separable metric space, the lookdown space, which we endow with a family of sampling measures. This yields a pathwise construction of tree-valued Fleming-Viot processes. In the case of coming down from infinity, we also read off a process whose state space is endowed with the Gromov-Hausdorff-Prohorov topology. This process has additional jumps at the extinction times of parts of the population.
In the case with only binary reproduction events, we construct the lookdown space also from the Aldous continuum random tree by removing the root and the highest leaf, and by deforming the metric in a way that corresponds to the time change that relates the Fleming-Viot process with a Dawson-Watanabe process. The sampling measures on the lookdown space are then image measures of the normalized local time measures.
We also show invariance principles for Markov chains that describe the evolving genealogy in Cannings models. For such Markov chains with values in the space of distance matrix distributions, we show convergence to tree-valued Fleming-Viot processes under the conditions of Möhle and Sagitov for the convergence of the genealogy at a fixed time to a coalescent with simultaneous multiple mergers. For the convergence of Markov chains with values in the space of marked metric measure spaces, an additional assumption is needed in the case with dust.
We document that natural disasters significantly weaken the stability of banks with business activities in affected regions, as reflected in lower z-scores, higher probabilities of default, higher non-performing assets ratios, higher foreclosure ratios, lower returns on assets and lower bank equity ratios. The effects are economically relevant and suggest that insurance payments and public aid programs do not sufficiently protect bank borrowers against financial difficulties. We also find that the adverse effects on bank stability dissolve after some years if no further disasters occur in the meantime.
This paper applies the theory of structured finance to the regulation of asset backed securities. We find the current regulation in Europe (Article 405 of the CRR) and the US (Section D of Dodd-Frank Act) to be severely flawed with respect to its key intention: the imposition of a strict loss retention requirement. While nominal retention is always 5%, the true level of loss retention varies across available retention options between zero loss retention and full loss retention at the extreme ends. Based on a standard model of structured finance transactions, we propose a new risk retention metric RM measuring the level of an issuer’s skin-in-the-game. The new metric could help to achieve a better implementation of CRR/CRD-IV and DFA, by making disclosure of the RM-number compulsory for all ABS transactions. There are also implications for the operation of rating agencies. On a general level, the RM metric will be instrumental in achieving simplicity and transparency in securitizations (STS).
Die erste Phase des im Dezember 2016 in Kraft getretenen Friedensabkommens zwischen der kolumbianischen Regierung und den FARC-Rebellen ist mit der Ankunft von ca. 6300 Kämpfern in den vereinbarten 26 Konzentrationszonen abgeschlossen. Dort wird nun der Prozess der Entwaffnung beginnen und die Vorbereitung auf die Eingliederung in das zivile Leben, ein Schritt, der der kolumbianischen Gesellschaft noch große Opfer abverlangen wird – sei es bezogen auf die Prozesse der justiziellen Aufarbeitung der Vergangenheit, sei es hinsichtlich der notwendigen Versöhnungsprozesse oder sei es bei der Suche nach neuen Formen des friedlichen Zusammenlebens. Sich in das zivile Leben einzufinden, dürfte insbesondere den ca. 7000 geschätzten Kindersoldaten Kolumbiens schwer fallen, die teilweise bereits im Alter von 12 Jahren von den illegalen bewaffneten Akteuren des Landes an Waffen ausgebildet wurden und ihre Kindheit bzw. Jugend in Guerilla-Verbänden verbracht haben. Die umfassende Betreuung der Kindersoldaten ist einer der zentralen Indikatoren für einen erfolgreichen Verlauf des Friedensprozesses in Kolumbien, wenn der Teufelskreis aus Gewalt, Vertreibung und Rekrutierung Minderjähriger durchbrochen werden soll. Sonst droht eine Verlängerung von Gewaltbiographien, die die Geschichte des Landes bereits in der Vergangenheit maßgeblich geprägt haben.
Am Donnerstag, dem 26.01.17, beschloss der Deutsche Bundestag, wie schon im letzten Jahr, eine Ausweitung des Bundeswehreinsatzes im Rahmen der UN-Mission MINUSMA in Mali. Die maximale Anzahl der in und um Gao im Norden Malis eingesetzten Soldatinnen und Soldaten wird von bisher 650 auf 1000 erhöht, da Deutschland zukünftig auch die Bereitstellung von Kampf- und Rettungshubschraubern für MINUSMA übernimmt. Damit wird der Einsatz in Mali zum Größten der Bundeswehr. Der Befehlshaber des Einsatzführungskommandos hat das Gefährdungspotenzial schon im vergangenen Jahr mit der ISAF-Mission in Afghanistan verglichen und in keinem anderen UN-Einsatz sind im letzten Jahr mehr Soldaten getötet worden.
Die Entsendung weiterer Soldatinnen und Soldaten vom Deutschen Bundestag in einen Einsatz, in dem sie erheblichen Gefahren für Leib und Leben ausgesetzt sind, erfordert eine umfangreiche Begründung durch die politisch Verantwortlichen und eine kritische Würdigung durch die Zivilgesellschaft.
Ist es zum Lachen oder zum Weinen? Diese Frage stellt sich immer wieder bei der Beobachtung der Worte und Taten des neuen US-Präsidenten – zum Beispiel anläßlich seines ersten Fernsehinterviews mit dem Journalisten David Muir (ABC America, 27.1.2017). Trump benahm sich wie ein rechthaberisches, selbstbezogenes, liebesbedürftiges Kind. Er beharrte auf seiner Version der Amtseinführung, nach der noch nie so viele Menschen wie diesmal an der Zeremonie teilgenommen hätten. Er sprach nicht nur davon, er verwies auch auf Fotos, die er an Wänden im Weißen Haus hat aufhängen lassen. Trump wiederholte außerdem seine Behauptung, dass es viele illegale Stimmen gegeben hätte, und alle für Hillary Clinton. Natürlich würde man auch den einen oder anderen finden, der illegal für ihn abgestimmt habe. Diese Person würde man dann, sagte Trump, als Gegenbeweis vor die Kameras zerren. Aber die Wahrheit sei, dass Millionen von illegalen Stimmen fast ausnahmslos für Clinton abgegeben worden seien...
Vom Wünschen

Es freut mich, dass ich bei der Gelegenheit meiner Antrittsvorlesung über ein Thema sprechen kann, das es mir schon lange angetan hat: das Wünschen. Ich werde gleich in einem einleitenden Abschnitt skizzieren, wie ich den Umfang und Inhalt des Themas bestimme und welche Fragen sich für mich daran anschließen. Hauptsächlich möchte ich mich dann auf einen Aspekt konzentrieren, der meiner Professur hier am Institut für Kulturwissenschaft der Humboldt-Universität zu Berlin besonders entspricht. Das Fachgebiet lautet "Kulturforschung mit Schwerpunkt Wissensgeschichte", und deshalb möchte ich in dieser Antrittsvorlesung vor allem über den Zusammenhang von Wünschen und Wissen sprechen, und somit auch über die Spannungen zwischen beidem. Es soll um die Frage gehen, was man vom Wünschen wissen kann, um die Frage, wie Wünschen das Wissen initiiert oder antreibt, aber auch darum, wie es das Wissen behindern oder sogar verhindern kann.
1777 veröffentlicht Lessing 'Über den Beweis des Geistes und der Kraft', eine seiner Streitschriften zur Bibel. Letztlich, argumentiert er hier, sei die Echtheit der Bibel gar nicht entscheidend, weil historische Tatsachen ohnehin nicht von allgemeinen Wahrheiten überzeugen könnten. Dieser Unterschied von zufälligen Geschichtswahrheiten und notwendigen Vernunftwahrheiten sei, so die berühmte Formulierung, der "garstige breite Graben" über den er nicht kommen könne, so oft und ernstlich er auch den Sprung versucht habe. So einen Graben mag einer vor sich sehen, wenn er vor einer Antrittsvorlesung steht, einem akademischen Schwellenritual, das wo keinen Sprung so doch einen entschiedenen Schritt erfordert. So einen Graben gibt es auch in der Sache: Man steht vor einem weiten Feld - in meinem Fall Kulturforschung mit Schwerpunkt Religion - dessen Grenzen sich im Ungefähren verlieren. Aber es ist nicht ganz leicht, auf dieses Feld zu kommen, es gibt Hindernisse, Schwellen, mögliche Missverständnisse - eben einen Graben. Es ist nicht mehr wie bei Lessing der Graben von Geschichte und Vernunft oder wie später bei Kierkegaard die Kluft zwischen Vernunft und Glauben. Eher schon ist es der historische Abstand, mit dem jede kulturgeschichtliche Untersuchung zu tun hat. In meinem Fall ist es auch noch ein anderer Graben, es ist der Abstand oder die Spannung von Kultur und Religion - von Forschung einmal zu schweigen -, und damit verbunden auch die Spannung von Religion und Moderne. Denn es ist nicht ganz leicht, Kultur und Religion in ein Verhältnis zu bringen, und es ist es besonders schwer in der Moderne.
Die hier veröffentlichten Vorträge wurden von Daniel Weidner und Stefan Willer am 9. Juni 2016 als Antrittsvorlesungen am Institut für Kulturwissenschaft der Humboldt-Universität zu Berlin gehalten. Daniel Weidner ist dort Professor für Kulturforschung mit dem Schwerpunkt Religion, Stefan Willer Professor für Kulturforschung mit dem Schwerpunkt Wissensgeschichte. Beide sind stellvertretende Direktoren des ZfL.
Jacob Taubes (1923-1987) und Carl Schmitt (1888-1985) zählen zu den umstrittenen, aber auch faszinierenden intellektuellen Figuren der Bundesrepublik: hier der jüdische Denker und Religionsphilosoph, ausgewiesener Kenner der apokalyptischen Strömungen in Judentum, Christentum, Gnosis samt ihren Folgen, die - nicht nur im christlichen Sinn - Filiationen innerhalb der Antike und ihres Nachlebens darstellen, dort der katholische Autor und Staatsrechtler, bekannt als Wortführer politischer Theologie, berüchtigt für seinen "aufhaltsamen Aufstieg zum 'Kronjuristen'" (Mehring) des Nationalsozialismus - ein Umstand, der sein Werk bis heute tief verschattet. Taubes und Schmitt haben polarisiert und tun es immer noch. Zu Lebzeiten provozierten sie durch ein Verhalten, das vielen als gemein, willkürlich, oft auch irritierend erschien, weil es übliche akademische Kabalen und Winkelzüge auf brüske Weise überschritt. Auch ergriffen sie rücksichtslos Partei, selbst bis zur fast völligen Isolierung: etwa wenn sich Schmitt 1933 als einziger aus der Kölner Juridischen Fakultät weigerte, für den Kollegen Hans Kelsen zu sprechen, als dieser von den Nationalsozialisten entlassen worden war (vgl. Rüthers), etwa wenn Taubes sich in erbitterte, persönliche Verletzungen bereitwillig in Kauf nehmende, akademische Kämpfe verstrickte, die er Ende der 1970er Jahre am notorischen Fachbereich 11 der Freien Universität Berlin [im Folgenden: FU] gegen nahezu alle führte. Doch verstanden es beide auch, geistig anzuziehen und intellektuell anzuregen, wenn sie Debatten begannen oder in sie eingriff en und Ideen aufnahmen, eben eine akademische Kardinaltugend aufs Beste beherrschten: das Verknüpfen von Wissen zwischen den Fächern, um persönliche oder sachliche Verbindungen für neue Fragen produktiv zu machen. Gerade durch die Missachtung gesetzter Grenzen ihrer jeweiligen Disziplin vermochten sie zu einer reicheren Erkenntnis vorzudringen. Darin sind sie Pioniere: Schmitt am Schnittpunkt von staatlichem Recht und christlicher Religion, Taubes in den Konstellationen jüdisch-christlicher Debatten jenseits aller konfessionell betriebenen Bemühungen um Dialog und Versöhnung. Weil ihr Einsatz als Denker - ungeachtet von Eskapaden und Eklats - ein geistig-existentieller war, wirken ihre Impulse weiter, in den letzten Jahren sogar zunehmend, davon zeugen die amerikanischen Übersetzungen, die viele ihrer Schriften und Texte erfahren haben, davon zeugt das Interesse, das sie gerade bei Jüngeren in Europa, Israel und den USA finden, die Philosophie, Jüdische Studien, Kultur-, Literatur-, Religionswissenschaft betreiben.
Positive Effekte körperliche Aktivität als komplementäre Therapie in der Onkologie wurden in den letzten Jahren in zahlreichen Studien aufgezeigt. Hierbei zeigte sich ein Anstieg der körperlichen Fitness und Muskelmasse, eine Steigerung der Lebensqualität, eine Reduktion des Fatigue-Syndroms, aber auch eine verbesserte Therapieverträglichkeit sowie einer Rezidiv-Prophylaxe (Backman et al., 2014; Meyerhardt et al., 2006; Segal et al., 2001). Daraufhin wurden Empfehlungen für körperliche Aktivität im Rahmen der onkologischen Therapie ausgesprochen, welche 150 Minuten moderate körperliche Aktivität pro Woche umfassen. Diese Empfehlungen basieren auf entitätsunspezifische Studienkollektive mit meist Tumorstadium I und II. Eine Vielzahl an Studien verdeutlichen, dass gerade Patienten in fortgeschrittenen Tumorstadien mit zahlreichen therapie- sowie tumorbedingten Nebenwirkungen zu kämpfen haben und dadurch ein stärkerer Abbau der körperlichen Fitness, der Muskulatur, aber auch der funktionellen Eigenschaften vorzufinden ist. Hierbei stellen Patienten mit fortgeschrittenen gastrointestinalen Tumoren (GIT) ein stark belastetes Kollektiv dar, da 80 Prozent dieser Patienten eine Tumorkachexie erleiden. Zusätzliche wurde in einer Querschnittsuntersuchung aufgezeigt, dass Patienten mit fortgeschrittenen GIT bereits vor Therapiestart einen deutlich verminderten körperlichen und funktionellen Status im Vergleich zu Mammakarzinom-Patientinnen und Gesunden aufweisen (Stuecher et al., 2016). In der vorliegenden randomisiert kontrollierten Untersuchung wurde erstmals ein heimbasiertes Training ohne Supervision zur Steigerung der körperlichen Aktivität bei Patienten mit fortgeschrittenen GIT durchgeführt und dieses mit einer leitliniengetreuen onkologischen Therapie ohne komplementäre Bewegungstherapie verglichen. Dabei wurden der körperliche und funktionelle Status sowie die Aktivitäten des täglichen Lebens verglichen.
Zweiundvierzig Patienten mit fortgeschrittenen GIT (UICC ≥ III, 67,1 ± 6,8 Jahre, 45,2 % weiblich) wurden vor ihrer geplanten first-line Chemotherapie (CT) in die zweiarmige randomisiert kontrollierte Studie eingeschlossen. Eine der Gruppen (I) erhielt, entsprechend der ACSM-Guidelines für onkologische Patienten, die Vorgabe ein wöchentliches Laufprogramm mit einem Umfang von 150 Minuten moderater Intensität pro Woche. Die zweite Gruppe diente als Kontrollgruppe (K) und erhielt am Ende der Studiendauer entsprechende Empfehlungen. Die Interventionsdurchführung wurde mittels Trainingstagebuch und Pedometer begleitet. Vor Beginn (T0), nach zwei CT-zyklen (T1) sowie nach zwölf Wochen (T2) wurde der funktionelle und körperliche Status sowie die Alltagsbewältigung der Patienten erfasst.
Bei einer Dropoutrate von 36 Prozent konnten 28 (K: 15; I; 13) Patienten die Studie komplett durchlaufen. Die mittlere Adhärenzrate lag bei 81,3 Prozent. Im Untersuchungszeitraum konnten die folgenden sign. Veränderungen (p< 0,05) der einzelnen Parameter gezeigt werden. Die posturale Stabilität (COPLänge) konnte sowohl ein Gruppeneffekt, als auch ein Zeiteffekt nachgewiesen werden. Die Interventionsgruppe verbesserte sich im Zeitraum T0-T1 (-71,47mm) sowie im Gesamtzeitraum T0-T2 (-74,13 mm), wohingegen die Kontrollgruppe sich im Gesamtzeitraum T0-T2 (+72,83) verschlechterte. Die Gruppen unterschieden sich daher sowohl in den Zeiträumen T0-T1 ((K)+38,61; (I)-71,47 mm) sowie T0-T2 ((K)+72,83; (I) -74,13mm). Bezüglich des körperlichen Status konnte sich die Interventionsgruppe von T1-T2 (+4,03 kg) sowie von T0-T2 (+4,04 kg) verbessern, sodass sich die Gruppen zwischen den Zeitpunkten T1-T2 ((K) -0,49; (I)+4,03 kg) und T0-T2 ((K) 0,19; 4,04 kg) unterschieden. Der iADL-Fragebogen erbrachte eine Verbesserung der Interventionsgruppe im Gesamtmesszeitraum T0-T2 (+0,12), daraus resultierte ein zusätzlicher Gruppenunterschied in diesem Zeitraum ((K) -0,89; (I) +0,12). Der Ernährungszustand zeigte auch einen unterschiedlichen Verlauf der beiden Gruppen. Zwischen T1-T2 ((K) -0,59; + 1,74) sowie T0-T2 ((K) -0,55; (I) +2,39) unterschieden sich die Gruppen.
Obgleich es für einige Patienten schwierig war die Laufintervention gemäß den Empfehlungen durchzuführen, weisen die Teilnehmer der Interventionsgruppe sowohl in den Parametern des körperlichen, als auch des funktionellen Status Verbesserungen auf. Demnach scheint ein durchschnittlicher Umfang von zwei Stunden moderater körperlicher Aktivität während einer Tumortherapie ausreichend zu sein. Es veranschaulicht, dass eine komplementäre Bewegungstherapie in der onkologischen Therapie bei Patienten mit fortgeschrittenen GIT sinnvoll ist und sowohl einen Benefit in der Körperzusammensetzung, als auch der funktionellen Eigenschaften mitsichbringt. Dies hat wiederum einen positiven Einfluss auf die Alltagsbewältigung. Da einige Barrieren das Laufprogramm der Patienten einschränkten oder gar zum Laufabbruch führten, sollte versucht werden diese zu mindern. Hierbei sind vor allem klima- und wetterbedingte Barrieren ein möglicher Ansatzpunkt, da Nebenwirkungen kaum vermeidbar sind. Dennoch sollten die Patienten auch nach nebenwirkungsbedingten Laufpausen motiviert werden das Programm weiterzuführen. Diese Studie gibt zudem erste Hinweise, dass durch eine komplementäre Bewegungstherapie mit moderater körperlicher Aktivität die Toxizität der CT bei Patienten mit fortgeschrittenen GIT vermindert werden kann. Da der klinische Benefit, welcher in einigen Studien anderer Tumorentitäten postuliert wurde, in dieser Untersuchung nicht objektiv erfasst wurde, wäre dies ein möglicher Ansatzpunkt für Folgestudien.
Ziel der Arbeit war es, die Flugzeitmassenspektrometrie als neue Analysemethode für die instrumentelle Analytik halogenierter Spurengase in der Luft zu etablieren. Die grundle-gende Motivation dafür ist, dass anthropogene Emissionen vieler Vertreter dieser Sub-stanzklasse einen negativen Einfluss auf die Umwelt zeigen: in der Atmosphäre agieren die Substanzen bzw. ihre Abbauprodukte als Katalysatoren für den stratosphärischen Ozonab-bau und verstärken den Strahlungsantrieb der Erde durch Absorption elektromagnetischer Strahlung im sogenannten atmosphärischen Fenster. Um diese Effekte und deren Auswir-kung quantifizieren zu können, ist es notwendig, Konzentrationen und Trends der Substan-zen in der Atmosphäre zu überwachen. Nur so können Gegenmaßnahmen wie Produktions-reglementierungen geplant und bewertet werden. In Kombination mit inverser Modellie-rung können zudem Rückschlüsse über tatsächlich emittierten Mengen gezogen werden. Dies stellt den Anspruch an die Analytik, sehr geringe Mengen dieser Gase sehr präzise quantifizieren zu können, um auch schwache Trends zu erkennen. Zudem muss die Analy-semethode die Möglichkeit zu bieten, mit der wachsenden Anzahl bekannter und zu über-wachender Substanzen Schritt zu halten. Besonders für letzteren Aspekt bietet die Flug-zeitmassenspektrometrie einen entscheidenden Vorteil gegenüber der „konventionellen“ Methode, der Quadrupolmassenspektrometrie: sie zeichnet das gesamten Massenspektrum auf ohne dadurch an Empfindlichkeit einzubüßen. Um das atmosphärische Mischungsver-hältnis von Substanzen im Bereich von pmol mol−1 bis fmol mol−1 bestimmen zu können, muss das Quadrupolmassenspektrometer im Single Ion Monitoring Modus betrieben wer-den – so wird zwar eine hohe Sensitivität erreicht, es wird aber auch nur die Intensität eines bestimmten Masse zu Ladungsverhältnisses (kurz: Masse) zu einem Zeitpunkt aufgezeich-net. Ein Flugzeitmassenspektrometer hingegen extrahiert Ionen mit einer Frequenz im Ki-loherzbereich und zeichnet für jede Extraktion das vollständige Flugzeitspektrum und da-mit Massenspektrum auf.
Aufgabe dieser Arbeit war es, ein Flugzeitmassenspektrometer mit vorgeschalteter Pro-benanreicherungseinheit sowie Gaschromatograph zur Trennung des Subtanzgemisches vor der Detektion aufzubauen und Werkzeuge zur Datenauswertung zu entwickeln. Um einen zukünftigen Feldeinsatz vorzubereiten, sollte der Aufbau möglichst kompakt, mobil und vollständig automatisiert sein. Anschließend sollte Empfindlichkeit, Präzision und dynami-scher Messbereich geprüft, optimiert und die Anwendbarkeit zur Analyse halogenierter Spurengase gezeigt werden. Die Ergebnisse aus der in der vorliegenden Arbeit präsentier-ten Geräteentwicklung finden sich in drei Publikationen wieder, welche in thematischer Reihenfolge die Probenanreicherung (Obersteiner et al., 2016b), den Vergleich von Quadrupol- und Flugzeitmassenspektrometrie (Hoker et al., 2015) sowie Eigenschaften und Anwendung des neuen Aufbaus (Obersteiner et al., 2016a) behandeln. Mit den genannten Aufsätzen ist die Arbeitsgruppe Engel weltweit die erste, welche hochpräzise Analytik ha-logenierter Spurengase routinemäßig mittels Flugzeitmassenspektrometrie durchführt. Der nächste Schritt ist der Übergang von der Laboranwendung zur Feldmessung, z.B. in Form von bodenbasierter in situ Analyse troposphärischer Luftmassen am Taunus Observatorium auf dem Kleinen Feldberg. Da es bisher keine Messstation für die hier beschriebene analy-tische Fragestellung in Deutschland gibt, könnte eine deutliche Verbesserung der Überwa-chung halogenierter Treibhausgase und ozonzerstörender Substanzen in Europa erzielt wer-den. Weiterhin wäre eine Flugzeugapplikation in Zukunft denkbar, welche neben der durch das Flugzeitmassenspektrometer abgedeckten Substanzbandbreite auch von dessen hoher möglicher Spektrenrate profitieren könnte. In Kombination mit Hochgeschwindigkeitsgas-chromatographie könnte eine bisher unerreichte Zeitauflösung der Beprobung der Atmo-sphäre mittels Gaschromatographie-Massenspektrometrie erzielt werden.
Der Einfluss des Klimas und Wetters auf den menschlichen Körper rückte seit dem frühen 19. Jahrhundert zunehmend in den Mittelpunkt wissenschaftlicher Untersuchungen. Die unmerkliche Allianz mit einer um die Jahrhundertwende populär werdenden naturheilpraktischen Diagnostik ließ einen ganzen Schwung meteorobiologischer Schriften folgen, und diese proklamierten einhellig den gegenseitigen Austausch zweier Systeme: des menschlichen Körpers und des ihn umgebenden Luftraums. Dabei handelte es sich zunächst um einen Raum ohne greifbare Gegenständlichkeit. Es ist demnach kein Zufall, dass eine vergleichbare Korrelationsbildung nun auch in der kunsttheoretischen Diskussion um den Einsatz der bildnerischen Mittel, jener "malerischen Ausdehnung des Raumes", wie Kandinskij es nannte, aktuell wurde.
Als im Sommer 1912 die zweite Optical Convention in London ihre Türen öffnete, füllten die ausgestellten Instrumente gleich mehrere Räume im Imperial College und im Science Museum in South Kensington. Alle einschlägigen Firmen waren nach London gekommen, um ihre Mikroskope, Kameras, Teleskope, Theodoliten, Spektroskope oder Ophthalmoskope einer interessierten Öffentlichkeit vorzustellen. Da bedurfte es schon einer raffinierten Inszenierung und einer gezielten Medienkampagne, um die Aufmerksamkeit des Publikums auch noch für das 'Optophon' zu mobilisieren, einen vergleichsweise unscheinbaren Holzkasten. Bereits am Vortag seiner öffentlichen Vorführung war deshalb mit entsprechend viel Fanfare in der Londoner 'Pall Mall Gazette' zu lesen: "To-morrow the Optical Convention is to let loose a new invention on the world. An ingenious Birmingham scientist has turned the element of selenium to account by making light audible, and we are to be dazzled and deafened both at once. Sunlight makes a roaring sound, and lightening, presumably, anticipates its concomitant thunder. All we require now is to increase the anticipative process, and then day light will awaken us every morning a couple of minutes before it arrives. What a point for the day light savers!"
The development of single-photon-counting detectors, such as the PILATUS, has been a major recent breakthrough in macromolecular crystallography, enabling noise-free detection and novel data-acquisition modes. The new EIGER detector features a pixel size of 75 × 75 µm, frame rates of up to 3000 Hz and a dead time as low as 3.8 µs. An EIGER 1M and EIGER 16M were tested on Swiss Light Source beamlines X10SA and X06SA for their application in macromolecular crystallography. The combination of fast frame rates and a very short dead time allows high-quality data acquisition in a shorter time. The ultrafine φ-slicing data-collection method is introduced and validated and its application in finding the optimal rotation angle, a suitable rotation speed and a sufficient X-ray dose are presented. An improvement of the data quality up to slicing at one tenth of the mosaicity has been observed, which is much finer than expected based on previous findings. The influence of key data-collection parameters on data quality is discussed.
Despite an increasing demand for Burgundy truffles (Tuber aestivum), gaps remain in our understanding of the fungus’ overall lifecycle and ecology. Here, we compile evidence from three independent surveys in Hungary and Switzerland. First, we measured the weight and maturity of 2,656 T. aestivum fruit bodies from a three-day harvest in August 2014 in a highly productive orchard in Hungary. All specimens ranging between 2 and 755 g were almost evenly distributed through five maturation classes. Then, we measured the weight and maturity of another 4,795 T. aestivum fruit bodies harvested on four occasions between June and October 2015 in the same truffière. Again, different maturation stages occurred at varying fruit body size and during the entire fruiting season. Finally, the predominantly unrelated weight and maturity of 81 T. aestivum fruit bodies from four fruiting seasons between 2010 and 2013 in Switzerland confirmed the Hungarian results. The spatiotemporal coexistence of 7,532 small-ripe and large-unripe T. aestivum, which accumulate to ~182 kg, differs from species-specific associations between the size and ripeness that have been reported for other mushrooms. Although size-independent truffle maturation stages may possibly relate to the perpetual belowground environment, the role of mycelial connectivity, soil property, microclimatology, as well as other abiotic factors and a combination thereof, is still unclear. Despite its massive sample size and proof of concept, this study, together with existing literature, suggests consideration of a wider ecological and biogeographical range, as well as the complex symbiotic fungus-host interaction, to further illuminate the hidden development of belowground truffle fruit bodies.
Pro-resolving lipid mediator Resolvin D1 serves as a marker of lung disease in cystic fibrosis

Background: Cystic fibrosis (CF) is an autosomal recessive genetic disorder that affects multiple organs, including the lungs, pancreas, liver and intestine. Mutations in the cystic fibrosis transmembrane conductance regulator (CFTR) locus lead to defective proteins and reduced Cl- secretion and Na+ hyperabsorption in the affected organs. In addition, patients suffering from CF display chronic inflammation that contributes to the pathogenesis of CF. Recent work suggests that CF patients have a reduced capacity to biosynthesize specialized pro-resolving lipid mediators (SPMs), which contributes to the development and duration of the unwanted inflammation. Alterations in the metabolism of arachidonic acid (AA) and docosahexaenoic acid (DHA) to specialized pro-resolving mediators (SPMs), like lipoxins (LXs), maresins (MaRs), protectins (PDs) and resolvins (Rvs), may play a major role on clinical impact of airway inflammation in CF.
Methods: In this study, our aims were to detect and quantitate Resolvin D1 (RvD1) in sputum and plasma from patients with CF and compare levels of RvD1 with biomarkers of inflammation and lung function. We studied 27 CF patients aged 6 to 55 years (median 16 years) in a prospective approach.
Results: DHA can be found in the plasma of our CF patients in the milligram range and is decreased in comparison to a healthy control group. The DHA-derived pro-resolving mediator Resolvin D1 (RvD1) was also present in the plasma (286.4 ± 50 pg/ mL, mean ± SEM) and sputum (30.0 ± 2.6 pg/ mL, mean ± SEM) samples from our patients with CF and showed a positive correlation with sputum inflammatory markers. The plasma concentrations of RvD1 were ten times higher than sputum concentrations. Interestingly, sputum RvD1/ IL-8 levels showed a positive correlation with FEV1 (rs = 0.3962, p< 0.05).
Conclusions: SPMs, like RvD1, are well known to down-regulate inflammatory pathways. Our study shows that the bioactive lipid mediator RvD1, derived from DHA, was present in sputum and plasma of CF patients and may serve as a representative peripheral biomarker of the lung resolution program for CF patients.
Objective: To investigate if plasma HIV-1 tropism testing could identify subjects at higher risk for clinical progression and death in routine clinical management.
Design: Nested case-control study within the EuroSIDA cohort.
Methods: Cases were subjects with AIDS or who died from any cause, with a plasma sample with HIV-1 RNA >1000 copies/mL available for tropism testing 3 to 12 months prior to the event. At least 1 control matched for age, HIV-1 RNA and HCV status at the time of sampling were selected per each case. Conditional logistic regression was used to investigate exposures associated with clinical progression to AIDS or death. A linear mixed model with random intercept was used to compare CD4+T-cell slopes by HIV tropism over the 12 months following the date of sampling.
Results: The study included 266 subjects, 100 cases and 166 controls; one quarter had X4 HIV; 26% were ART-naïve. Baseline factors independently associated with clinical progression or death were female gender (OR = 2.13 vs. male, 95CI = 1.04, 4.36), p = 0.038), CD4+T-cell count (OR = 0.90 (95CI = 0.80, 1.00) per 100 cells/mm3 higher, p = 0.058), being on ART (OR = 2.72 vs. being off-ART (95CI = 1.15, 6.41), p = 0.022) and calendar year of sample [OR = 0.84 (95CI = 0.77, 0.91) per more recent year, p<0.001). Baseline tropism was not associated with the risk of clinical progression or death. CD4+T-cell slopes did not differ within or between tropism groups.
Conclusions: The predictive role of plasma tropism determined using 454 sequencing in the context of people receiving cART with detectable VL is not helpful to identify subjects at higher risk for clinical progression to AIDS or death.
Cannabinoid antagonist SLV326 induces convulsive seizures and changes in the interictal EEG in rats

Cannabinoid CB1 antagonists have been investigated for possible treatment of e.g. obesity-related disorders. However, clinical application was halted due to their symptoms of anxiety and depression. In addition to these adverse effects, we have shown earlier that chronic treatment with the CB1 antagonist rimonabant may induce EEG-confirmed convulsive seizures. In a regulatory repeat-dose toxicity study violent episodes of “muscle spasms” were observed in Wistar rats, daily dosed with the CB1 receptor antagonist SLV326 during 5 months. The aim of the present follow-up study was to investigate whether these violent movements were of an epileptic origin. In selected SLV326-treated and control animals, EEG and behavior were monitored for 24 hours. 25% of SLV326 treated animals showed 1 to 21 EEG-confirmed generalized convulsive seizures, whereas controls were seizure-free. The behavioral seizures were typical for a limbic origin. Moreover, interictal spikes were found in 38% of treated animals. The frequency spectrum of the interictal EEG of the treated rats showed a lower theta peak frequency, as well as lower gamma power compared to the controls. These frequency changes were state-dependent: they were only found during high locomotor activity. It is concluded that long term blockade of the endogenous cannabinoid system can provoke limbic seizures in otherwise healthy rats. Additionally, SLV326 alters the frequency spectrum of the EEG when rats are highly active, suggesting effects on complex behavior and cognition.

In the title hydrate, C28H42N2O3·H2O, the central 1,3-diazinan-5-ol ring adopts a chair conformation with the two benzyl substituents equatorial and the lone pairs of the N atoms axial. The dihedral angle between the aromatic rings is 19.68 (38)°. There are two intramolecular O-H...N hydrogen bonds, each generating an S(6) ring motif. In the crystal, classical O-H...O hydrogen bonds connect the 1,3-diazinane and water molecules into columns extending along the b axis. The crystal structure was refined as a two-component twin with a fractional contribution to the minor domain of 0.0922 (18).
The title fluorinated bisbenzoxazine, C18H18F2N2O2, crystallizes with one half-molecule in the asymmetric unit, which is completed by inversion symmetry. The fused oxazine ring adopts an approximately half-chair conformation. The two benzoxazine rings are oriented anti to one another around the central C-C bond. The dominant intermolecular interaction in the crystal structure is a C-H...F hydrogen bond between the F atoms and the axial H atoms of the OCH2N methylene group in the oxazine rings of neighbouring molecules. C-H...[pi] contacts further stabilize the crystal packing.
In the title compound, C7H14N4·2C6H5ClO, which crystallized with two crystallographically independent 4-chlorophenol molecules and one 1,3,6,8-tetraazatricyclo[4.3.1.13,8]undecane (TATU) molecule in the asymmetric unit, the independent components are linked by two O-H...N hydrogen bonds. The hydrogen-bond acceptor sites are two non-equivalent N atoms from the aminal cage structure, and the tricyclic system distorts by changing the C-N bond lengths. In the crystal, these hydrogen-bonded aggregates are linked into chains along the c axis by C-H...N hydrogen bonds. The crystal structure also features C-H...[pi] contacts.
The title benzoxazine molecule, C18H18Br2N2O2, was prepared by a Mannich-type reaction of 4-bromophenol with ethane-1,2-diamine and formaldehyde. The title compound crystallizes in the monoclinic space group C2/c with a centre of inversion located at the mid-point of the C-C bond of the central CH2CH2 spacer. The oxazinic ring adopts a half-chair conformation. The structure is compared to those of other functionalized benzoxazines synthesized in our laboratory. In the crystal, weak C-H...Br and C-H...O hydrogen bonds stack the molecules along the b-axis direction.
Solvent-free treatment of 1,3,6,8-tetraazatricyclo[4.3.1.13,8]undecano (TATU) with 4-chloro-3,5-dimethylphenol led to the formation of the title co-crystal, C7H14N4·2C8H9ClO. The asymmetric unit contains one aminal cage molecule and two phenol molecules linked via two O-H...N hydrogen bonds. In the aminal cage, the N-CH2-CH2-N unit is slightly distorted from a syn periplanar geometry. Aromatic [pi]-[pi] stacking between the benzene rings from two different neighbouring phenol molecules [centroid-centroid distance = 4.0570 (11) Å] consolidates the crystal packing.
The formation of acquired drug resistance is a major reason for the failure of anti-cancer therapies after initial response. Here, we introduce a novel model of acquired oxaliplatin resistance, a sub-line of the non-MYCN-amplified neuroblastoma cell line SK-N-AS that was adapted to growth in the presence of 4000 ng/mL oxaliplatin (SK-N-ASrOXALI4000). SK-N-ASrOXALI4000 cells displayed enhanced chromosomal aberrations compared to SK-N-AS, as indicated by 24-chromosome fluorescence in situ hybridisation. Moreover, SK-N-ASrOXALI4000 cells were resistant not only to oxaliplatin but also to the two other commonly used anti-cancer platinum agents cisplatin and carboplatin. SK-N-ASrOXALI4000 cells exhibited a stable resistance phenotype that was not affected by culturing the cells for 10 weeks in the absence of oxaliplatin. Interestingly, SK-N-ASrOXALI4000 cells showed no cross resistance to gemcitabine and increased sensitivity to doxorubicin and UVC radiation, alternative treatments that like platinum drugs target DNA integrity. Notably, UVC-induced DNA damage is thought to be predominantly repaired by nucleotide excision repair and nucleotide excision repair has been described as the main oxaliplatin-induced DNA damage repair system. SK-N-ASrOXALI4000 cells were also more sensitive to lysis by influenza A virus, a candidate for oncolytic therapy, than SK-N-AS cells. In conclusion, we introduce a novel oxaliplatin resistance model. The oxaliplatin resistance mechanisms in SK-N-ASrOXALI4000 cells appear to be complex and not to directly depend on enhanced DNA repair capacity. Models of oxaliplatin resistance are of particular relevance since research on platinum drugs has so far predominantly focused on cisplatin and carboplatin.
Ongoing liver inflammation in patients with chronic hepatitis C and sustained virological response

Background: Novel direct-acting antiviral DAA combination therapies tremendously improved sustained virologic response (SVR) rates in patients with chronic HCV infection. SVR is typically accompanied by normalization of liver enzymes, however, hepatic inflammation, i.e. persistently elevated aminotransferase levels may persist despite HCV eradication. Aim: To investigate prevalence and risk factors for ongoing hepatic inflammation after SVR in two large patient cohorts.
Methods: This post-hoc analysis was based on prospectively collected demographic and clinical data from 834 patients with SVR after HCV treatment with either PegIFN- or DAA-based treatment regimens from the PRAMA trial (n = 341) or patients treated at our outpatient clinic (n = 493).
Results: We observed an unexpected high prevalence of post-SVR inflammation, including patients who received novel IFN-free DAA-based therapies. Up to 10% of patients had ongoing elevation of aminotransferase levels and another 25% showed aminotransferase activity above the so-called healthy range. Several baseline factors were independently associated with post-SVR aminotransferase elevation. Among those, particularly male gender, advanced liver disease and markers for liver steatosis were strongly predictive for persistent ALT elevation. The use of IFN-based antiviral treatment was independently correlated with post-SVR inflammation, further supporting the overall benefit of IFN-free combination regimens.
Conclusion: This is the first comprehensive study on a large patient cohort investigating the prevalence and risk factors for ongoing liver inflammation after eradication of HCV. Our data show a high proportion of patients with ongoing hepatic inflammation despite HCV eradication with potential implications for the management of approximately one third of all patients upon SVR.
The objective of this study was to characterize blaOXA-23 harbouring Acinetobacter indicus-like strains from cattle including genomic and phylogenetic analyses, antimicrobial susceptibility testing and evaluation of pathogenicity in vitro and in vivo. Nasal and rectal swabs (n = 45) from cattle in Germany were screened for carbapenem-non-susceptible Acinetobacter spp. Thereby, two carbapenem resistant Acinetobacter spp. from the nasal cavities of two calves could be isolated. MALDI-TOF mass spectrometry and 16S rDNA sequencing identified these isolates as A. indicus-like. A phylogenetic tree based on partial rpoB sequences indicated closest relation of the two bovine isolates to the A. indicus type strain A648T and human clinical A. indicus isolates, while whole genome comparison revealed considerable intraspecies diversity. High mimimum inhibitory concentrations were observed for carbapenems and other antibiotics including fluoroquinolones and gentamicin. Whole genome sequencing and PCR mapping revealed that both isolates harboured blaOXA-23 localized on the chromosome and surrounded by interrupted Tn2008 transposon structures. Since the pathogenic potential of A. indicus is unknown, pathogenicity was assessed employing the Galleria (G.) mellonella infection model and an in vitro cytotoxicity assay using A549 human lung epithelial cells. Pathogenicity in vivo (G. mellonella killing assay) and in vitro (cytotoxicity assay) of the two A. indicus-like isolates was lower compared to A. baumannii ATCC 17978 and similar to A. lwoffii ATCC 15309. The reduced pathogenicity of A. indicus compared to A. baumannii correlated with the absence of important virulence genes encoding like phospholipase C1+C2, acinetobactin outer membrane protein BauA, RND-type efflux system proteins AdeRS and AdeAB or the trimeric autotransporter adhesin Ata. The emergence of carbapenem-resistant A. indicus-like strains from cattle carrying blaOXA-23 on transposable elements and revealing genetic relatedness to isolates from human clinical sources requires further investigations regarding the pathogenic potential, genomic characteristics, zoonotic risk and putative additional sources of this new Acinetobacter species.
Bipolar disorder (BD) is a highly heritable neuropsychiatric disease characterized by recurrent episodes of mania and depression. BD shows substantial clinical and genetic overlap with other psychiatric disorders, in particular schizophrenia (SCZ). The genes underlying this etiological overlap remain largely unknown. A recent SCZ genome wide association study (GWAS) by the Psychiatric Genomics Consortium identified 128 independent genome-wide significant single nucleotide polymorphisms (SNPs). The present study investigated whether these SCZ-associated SNPs also contribute to BD development through the performance of association testing in a large BD GWAS dataset (9747 patients, 14278 controls). After re-imputation and correction for sample overlap, 22 of 107 investigated SCZ SNPs showed nominal association with BD. The number of shared SCZ-BD SNPs was significantly higher than expected (p = 1.46x10-8). This provides further evidence that SCZ-associated loci contribute to the development of BD. Two SNPs remained significant after Bonferroni correction. The most strongly associated SNP was located near TRANK1, which is a reported genome-wide significant risk gene for BD. Pathway analyses for all shared SCZ-BD SNPs revealed 25 nominally enriched gene-sets, which showed partial overlap in terms of the underlying genes. The enriched gene-sets included calcium- and glutamate signaling, neuropathic pain signaling in dorsal horn neurons, and calmodulin binding. The present data provide further insights into shared risk loci and disease-associated pathways for BD and SCZ. This may suggest new research directions for the treatment and prevention of these two major psychiatric disorders.
Historisch betrachtet wurde Krebspatienten durch das behandelnde Personal von physischer Belastungen oftmals abgeraten (Steins Bisschop et al., 2012). Mit der zunehmenden Zahl an Publikationen im Bereich onkologischer Rehabilitation wird deutlich, dass Krebspatienten oftmals eine stark reduzierte kardiorespiratorische Leistungsfähigkeit aufweisen, was wiederum Implikationen für akute wie auch längerfristige krebs-assoziierte Effekte und Outcomes haben kann (Jones et al., 2008). Somit steht dem bisherigen Verhaltens-Dogma der Ruhe und Schonung bei Krebspatienten eine Vielzahl an Evidenz der Effektivität und Notwendigkeit von regelmäßiger körperlicher Bewegung gegenüber (Jones et al., 2008; Klika et al., 2009; Steins Bisschop et al., 2012). Diese Arbeiten bieten fundierte Informationen bezüglich der zu Grunde liegenden Limitationen der Belastungstoleranz. Darauf aufbauend wird dieses Wissen genutzt, um effektive Trainings- und Rehabilitationsprogramme zu entwerfen sowie klinische Resultate zu verbessern (Jones et al., 2009).
Strukturierte und individualisierte Sport- und Bewegungstherapie gilt dabei als ein vielversprechender Ansatz, die nachlassende Fähigkeit der selbständigen Alltagsbewältigung, die krebsassoziierte Müdigkeitssymptomatik (fatigue), aber auch die allgemeine kardiorespiratorische Fitness (KRF) zu verbessern (Schmitz et al., 2010). Für onkologische Patienten von ebenso großer Bedeutung ist die Tatsache, dass durch regelmäßige körperliche Aktivität Begleiterscheinungen der eingesetzten Behandlungsmethoden (Hayes et al., 2009), das psychische Wohlbefinden als auch die Gesamtmortalität positiv beeinflusst werden können (Mishra et al., 2012b; Mishra et al., 2012a). Zusätzlich verspricht regelmäßige körperliche Bewegung, den Funktionsverlust in den drei wichtigsten physiologischen Bereichen (Muskulatur, Herz-Kreislauf-System und kardiopulmonales System) zu verringern oder sogar aufzuhalten (Hayes et al., 2009). Demzufolge nimmt die sporttherapeutische Sekundär- und Tertiärprävention bei Krebserkrankungen eine wichtige Rolle ein, und die Frage nach einer effektiven und zielgerichteten Belastungs- und Trainingssteuerung auf Basis valider und reliabler Messgrößen der körperlichen Leistungsfähigkeit gewinnt zunehmend an Bedeutung (Ulrich et al., 2013; Kirkham et al., 2013).
Aktuelle Empfehlungen zu körperlicher Aktivität bei onkologischen Patienten orientieren sich hinsichtlich der Ausgestaltung in punkto Häufigkeit, Umfang und Intensität im Allgemeinen an Bewegungs-Leitlinien für gesunde Gleichaltrige ohne chronische Erkrankungen. Eine Besonderheit bei onkologischen Patienten stellt jedoch oftmals die Berücksichtigung therapieassoziierter bzw. krankheitsbedingter Nebenwirkungen in der praktischen Umsetzung der Empfehlungen dar. Aktuelle Leitlinien sehen dafür ein aerobes Ausdauertraining an mindestens 3-5 Tagen/Woche bei einem Intensitätsspektrum moderater (150min) oder intensiver (75min) Beanspruchung (MVPA) bei 46-90% der maximalen Sauerstoffaufnahmefähigkeit vor.
Die in der Literatur beschriebenen Probleme einer Vielzahl an Verfahren zur Bestimmung der KRF und der darauf basierenden Ableitung von individualisierten Trainingsempfehlungen haben in den letzten Jahren vermehrt zu Ansätzen basierend auf der individuell physiologischen Belastungsreaktion in der Ausdauertrainingssteuerung und –bewertung geführt (Meyer et al., 2005c; Bentley et al., 2007b; Binder et al., 2008; Hofmann & Tschakert, 2011). Dabei kommen vor allem die in der sportmedizinischen Ausdauerleistungsdiagnostik etablierten Verfahren der Laktat-Leistungsdiagnostik und der spiroergometrischen Ermittlung ventilatorischer Schwellen zum Einsatz.
Kernziel der vorliegenden Arbeit war daher die Untersuchung von Leistungskennwerten an submaximalen Variablen aus Spiroergometrie und der Laktatdiagnostik im Kollektiv der onkologischen Patienten mit unterschiedlichen Krebsentitäten.
Das folgende Kapitel 2 fasst den aktuellen Forschungsstand zu Epidemiologie, Prävalenz sowie die Auswirkungen onkologischer Erkrankungen auf Aspekte der körperlichen Ausdauerleistungsfähigkeit und deren Diagnostik in der Sportmedizin zusammen. Resultierend aus dem Forschungsstand werden daraufhin Zielstellung und –fragen der vorliegenden Arbeit formuliert. Kapitel 4 umfasst die methodische Ablaufbeschreibung des Studiendesigns, die Probandenrekrutierung und -stichprobe, die eingesetzten diagnostischen Messverfahren, sowie die Darstellung der statistischen Auswertung. Nachfolgend auf die Darstellung der Ergebnisse in Kapitel 5 werden diese in Kapitel 6 vor dem Hintergrund des dargestellten Forschungstandes und aktueller relevanter Literatur diskutiert. Abschließend erfolgt eine Praxis-orientierte kritische Beleuchtung der Arbeit sowie ein eine perspektivische Einordnung der Ergebnisse mit Ausblick auf zukünftige Forschungsarbeiten.
In Germany, orthopedic workforce planning relies on population-to-provider-ratios represented by the "official degree of care provision". However, with geographic information systems (GIS), more sophisticated measurements are available. By utilizing GIS-based technologies we analyzed the current state of demand and supply of the orthopedic workforce in Germany (orthopedic accessibility) with the integrated Floating Catchment Area method. The analysis of n = 153,352,220 distances revealed significant geographical variations on national scale: 5,617,595 people (6.9% of total population) lived in an area with significant low orthopedic accessibility (average z-score = -4.0), whereas 31,748,161 people (39.0% of total population) lived in an area with significant high orthopedic accessibility (average z-score = 8.0). Accessibility was positively correlated with the degree of urbanization (r = 0.49; p<0.001) and the official degree of care provision (r = 0.33; p<0.001) and negatively correlated with regional social deprivation (r = -0.47; p<0.001). Despite advantages of simpler measures regarding implementation and acceptance in health policy, more sophisticated measures of accessibility have the potential to reduce costs as well as improve health care. With this study, significant geographical variations were revealed that show the need to reduce oversupply in less deprived urban areas in order to enable adequate care in more deprived rural areas.
Background: Previous magnetic resonance imaging (MRI) research suggests that, prior to the onset of psychosis, high risk youths already exhibit brain abnormalities similar to those present in patients with schizophrenia.
Objectives: The goal of the present study was to describe the functional organization of endogenous activation in young adolescents who report auditory verbal hallucinations (AVH) in view of the “distributed network” hypothesis of psychosis. We recruited 20 young people aged 13–16 years who reported AVHs and 20 healthy controls matched for age, gender and handedness from local schools.
Methods: Each participant underwent a semi-structured clinical interview and a resting state (RS) neuroimaging protocol. We explored functional connectivity (FC) involving three different networks: 1) default mode network (DMN) 2) salience network (SN) and 3) central executive network (CEN). In line with previous findings on the role of the auditory cortex in AVHs as reported by young adolescents, we also investigated FC anomalies involving both the primary and secondary auditory cortices (A1 and A2, respectively).
Further, we explored between-group inter-hemispheric FC differences (laterality) for both A1 and A2. Compared to the healthy control group, the AVH group exhibited FC differences in all three networks investigated. Moreover, FC anomalies were found in a neural network including both A1 and A2. The laterality analysis revealed no between-group, inter-hemispheric differences.
Conclusions: The present study suggests that young adolescents with subclinical psychotic symptoms exhibit functional connectivity anomalies directly and indirectly involving the DMN, SN, CEN and also a neural network including both primary and secondary auditory cortical regions.
