Visuals can be effective tools for educating an audience about peacebuilding and the need to engage with a nation's violent past. However, research on visuality has pointed to the ambivalence visuals can develop through audiencing and the dominant political discourse. Building on this, this article argues that ambivalence can also occur between narratives by different media although the same institution produced them, and that such inherent contradictions can limit the institution's effectiveness. The analysis centers upon a case study of the East Timorese Commission for Reception, Truth and Reconciliation (CAVR) that compares the commission's documentary dalan ba dame (“road to peace”) with its final report about peace and the human rights violations committed in the territory between 1975 and 1999. While the commission's final report stresses the individual responsibility of members of the Indonesian military and formulates the need for an institution-based liberal peace, the documentary communicates the message that all parties to the conflict are guilty of committing crimes and that peace has already been created, mitigating the need to further engage with the violent past. The analysis identifies the media's different formats and their different agendas as reasons for the creation of these contradicting messages. Based on an assessment of the dissemination of both media and their reception within the political discourse in Timor-Leste, the implications of these conflicting narratives for educating an international audience are discussed. Since the final report is difficult to access due to its length and its legal language, the documentary remains the more accessible medium to educate an international audience about the nation's violent past. However, due to the narrative it conveys, the documentary's ability to mobilize an international audience is limited. Thus, the article argues for considering three aspects when designing visuals for peace education: the intermediality of visuals with other media and its potential effects concerning the communication of a specific message, the reception of the message by the target audience, and the reception of the message by broader audiences when the visual is distributed online.
Lysine-specific demethylase 1 (LSD1), a histone lysine demethylase with the main specificity for H3K4me2, has been shown to be overexpressed in rhabdomyosarcoma (RMS) tumor samples. However, its role in RMS biology is not yet well understood. Here, we identified a new role of LSD1 in regulating adhesion of RMS cells. Genetic knockdown of LSD1 profoundly suppressed clonogenic growth in a panel of RMS cell lines, whereas LSD1 proved to be largely dispensable for regulating cell death and short-term survival. Combined RNA and ChIP-sequencing performed to analyze RNA expression and histone methylation at promoter regions revealed a gene set enrichment for adhesion-associated terms upon LSD1 knockdown. Consistently, LSD1 knockdown significantly reduced adhesion to untreated surfaces. Importantly, precoating of the plates with the adhesives collagen I or fibronectin rescued this reduced adhesion of LSD1 knockdown cells back to levels of control cells. Using KEGG pathway analysis, we identified 17 differentially expressed genes (DEGs) in LSD1 knockdown cells related to adhesion processes, which were validated by qRT-PCR. Combining RNA and ChIP-sequencing results revealed that, within this set of genes, SPP1, C3AR1, ITGA10 and SERPINE1 also exhibited increased H3K4me2 levels at their promoter regions in LSD1 knockdown compared to control cells. Indeed, LSD1 ChIP experiments confirmed enrichment of LSD1 at their promoter regions, suggesting a direct transcriptional regulation by LSD1. By identifying a new role of LSD1 in the modulation of cell adhesion and clonogenic growth of RMS cells, these findings highlight the importance of LSD1 in RMS.
The Asplenium coenobiale complex is distributed in Eastern Asia and Southeast Asia with its distribution center in southwestern China. In this study, we carried out a detailed morphological, cytological, and phylogenetic study by adding two samples from Danxia landform in Guangdong. The sequences of five chloroplast markers and one nuclear marker of the A. coenobiale complex were analyzed with maximum likelihood and Bayesian inference, respectively. The morphological and phylogenetic analyses support the recognition of a new species (A. danxiaense K.W.Xu sp. nov.) of the A. coenobiale complex from a cave of Danxia mountain, Guangdong province, southern China. This new species can be distinguished from A. coenobiale and A. pulcherrimum by having scales narrowly triangular to lanceolate, apex ending in a short apical tail, basal basiscopic pinnule usually largest, fertile segment scarce, and exospore length usually more than 50 μm and shows significant molecular differences from other species in this complex. A detailed description and illustrations are presented.
We describe three new species of the genus Coletinia Wygodzinsky, 1980, the most diverse of the family Nicoletiidae (order Zygentoma = Thysanura s. str.) in Europe: C. dalmatica Molero-Baltanás, Fišer, Bach de Roca & Gaju-Ricart sp. nov. from Croatia, C. dextra Molero-Baltanás, Bach de Roca & Gaju-Ricart sp. nov. from Spain and C. serrata Mendes, Molero-Baltanás, Bach de Roca & Gaju-Ricart sp. nov. from Portugal. Coletinia dalmatica sp. nov. seems to be related with C. maggi (Grassi, 1887) and both Iberian species belong to the asymetrica species group. New material of some previously recognized species (C. diania Molero, Bach & Gaju, 2013, C. intermedia Molero, Bach & Gaju, 2013, C. mendesi Wygodzinsky, 1980, C. redetecta Molero, Bach & Gaju, 2013 and C. tinauti Molero-Baltanás, Gaju-Ricart & Bach de Roca, 1997) is reported, enlarging in most cases their geographic distribution, and including some new data on their intraspecific variability. Finally, an updated identification key to all known species of the genus Coletinia is given.

First as a student of comparative literature with a focus on German and then as a professor of German Studies, I’ve been traveling back and forth to Germany for three decades, almost exactly the age of the reunified German state. I have stayed for weeks, for months, or for more than a year at a time. I have lived in Leipzig, in Cologne, and in Munich, but I have spent by far the most time in Berlin, a place that I have come to consider a second home. Throughout that time, Germany has changed enormously, both demographically and attitudinally. In relation to diversity in general and in its relationship to Jews.

Understanding hominin expansions requires the comprehension of movement processes at different scales. In many models of hominin expansion these processes are viewed as being determined by large-scale effects, such as changes in climate and vegetation spanning continents and thousands or even millions of years. However, these large-scale patterns of expansions also need to be considered as possibly resulting from the accumulation of small-scale decisions of individual hominins. Moving on a continental scale may for instance involve crossing a water barrier. We present a generalized agent-based model for simulating the crossing of a water barrier where the agents represent the hominin individuals. The model can be configured to represent a variety of movement modes across water. Here, we compare four different behavioral scenarios in conjunction with a set of water barrier configurations, in which agents move in water by either paddling, drifting, swimming or rafting. We introduce the crossing-success-rate (CSR) to quantify the performance in water crossing. Our study suggests that more focus should be directed towards the exploration of behavioral models for hominins, as directionality may be a more powerful factor for crossing a barrier than environmental opportunities alone. A prerequisite for this is to perceive the opposite shore. Furthermore, to provide a comprehensive understanding of hominin expansions, the CSR allows for the integration of results obtained from small-scale simulations into large-scale models for hominin expansion.
Abstract: The human visual cortex enables visual perception through a cascade of hierarchical computations in cortical regions with distinct functionalities. Here, we introduce an AI-driven approach to discover the functional mapping of the visual cortex. We related human brain responses to scene images measured with functional MRI (fMRI) systematically to a diverse set of deep neural networks (DNNs) optimized to perform different scene perception tasks. We found a structured mapping between DNN tasks and brain regions along the ventral and dorsal visual streams. Low-level visual tasks mapped onto early brain regions, 3-dimensional scene perception tasks mapped onto the dorsal stream, and semantic tasks mapped onto the ventral stream. This mapping was of high fidelity, with more than 60% of the explainable variance in nine key regions being explained. Together, our results provide a novel functional mapping of the human visual cortex and demonstrate the power of the computational approach.
Author Summary: Human visual perception is a complex cognitive feat known to be mediated by distinct cortical regions of the brain. However, the exact function of these regions remains unknown, and thus it remains unclear how those regions together orchestrate visual perception. Here, we apply an AI-driven brain mapping approach to reveal visual brain function. This approach integrates multiple artificial deep neural networks trained on a diverse set of functions with functional recordings of the whole human brain. Our results reveal a systematic tiling of visual cortex by mapping regions to particular functions of the deep networks. Together this constitutes a comprehensive account of the functions of the distinct cortical regions of the brain that mediate human visual perception.
Childbirth-related post-traumatic stress disorder (CB-PTSD) occurs in 3–7% of all pregnancies and about 35% of women after preterm birth (PTB) meet the criteria for acute stress reaction. Known risk factors are trait anxiety and pain intensity, whereas planned delivery mode, medical support, and positive childbirth experience are protective factors. It has not yet been investigated whether the effects of anxiety and delivery mode are mediated by other factors, and whether a PTB-risk alters these relationships. 284 women were investigated antepartum and six weeks postpartum (risk-group with preterm birth (RG-PB) N = 95, risk-group with term birth (RG-TB) N = 99, and control group (CG) N = 90). CB-PTSD symptoms and anxiety were measured using standardized psychological questionnaires. Pain intensity, medical support, and childbirth experience were assessed by single items. Delivery modes were subdivided into planned vs. unplanned delivery modes. Group differences were examined using MANOVA. To examine direct and indirect effects on CB-PTSD symptoms, a multi-sample path analysis was performed. Rates of PTS were highest in the RG-PB = 11.58% (RG-TB = 7.01%, CG = 1.1%). MANOVA revealed higher values of CB-PTSD symptoms and pain intensity in RG-PB compared to RG-TB and CG. Women with planned delivery mode reported a more positive birth experience. Path modeling revealed a good model fit. Explained variance was highest in RG-PB (R2 = 44.7%). Direct enhancing effects of trait anxiety and indirect reducing effects of planned delivery mode on CB-PTSD symptoms were observed in all groups. In both risk groups, CB-PTSD symptoms were indirectly reduced via support by medical staff and positive childbirth experience, while trait anxiety indirectly enhanced CB-PTSD symptoms via pain intensity in the CG. Especially in the RG-PB, a positive birth experience serves as protective factor against CB-PTSD symptoms. Therefore, our data highlights the importance of involving patients in the decision process even under stressful birth conditions and the need for psychological support antepartum, mainly in patients with PTB-risk and anxious traits.
Development and preliminary validation of the Emotions while Learning an Instrument Scale (ELIS)

Learning to play a musical instrument is associated with different, partially conflicting emotions. This paper describes the development and psychometric properties of the Emotions while Learning an Instrument Scale (ELIS). In a longitudinal study with 545 German elementary school children factorial structure and psychometric properties were evaluated. Exploratory and confirmatory factor analyses confirmed a two-factor solution measuring Positive musical Emotions while Learning an Instrument (PELI) and Negative Emotions while Learning an Instrument (NELI). Both subscales yielded scores with adequate internal reliability (Cronbach’s α = .74, .86) and relatively stable retest reliabilities over 18 months (r = .11 -.56). Preliminary evidence of congruent and divergent validity of the subscales is provided. Implications for future research of musical emotional experiences in children are discussed.
Introduction: Cesarean section (CS) rates are increasing worldwide. One constant indication is the breech presentation at term. By offering external cephalic version (ECV) and vaginal breech delivery CS rates can be further reduced. Objective: This study aimed to analyze the ECV at 38 weeks of gestation with the associate uptake rate, predicting factors, success rate, and complications at a tertiary healthcare provider in Germany specializing in vaginal breech delivery. Methods: We conducted a prospective cohort study with retrospective data acquisition. All women with a singleton fetus in breech presentation presenting after 34 weeks of gestation for counseling between 2013 and 2017 were included. ECV impact factors were analyzed using logistic regression. Results: A total of 1,598 women presented for breech birth planning. ECV was performed on 353 patients. The overall success rate was 22.4%. A later week of gestation (odds ratio [OR] 1.69), an abundant amniotic fluid index (AFI score) (OR 5.74), fundal (OR 3.78) and anterior (OR 0.39) placental location, and an oblique lie (OR 9.08) were significantly associated with successful ECV in our population. No major complications were observed. The overall vaginal delivery rates could be increased to approximately 14% with ECV. Conclusion: The demand for alternative birth modes other than CS for breech birth is high in the area of Frankfurt, Germany. Our study offers evidence of the safety of ECV at 38 weeks. Centers with expertise in vaginal breech delivery and ECV can reduce CS-rates. To further establish vaginal breech delivery and ECV as alternate options, the required knowledge and skill should be implemented in the revised curricula.
Background: Pathogenesis of portal hypertension is multifactorial and includes pathologic intrahepatic angiogenesis, whereby TIPS insertion is an effective therapy of portal hypertension associated complications. While angiogenin is a potent contributor to angiogenesis in general, little is known about its impact on TIPS function over time. Methods: In a total of 118 samples from 47 patients, angiogenin concentrations were measured in portal and inferior caval vein plasma at TIPS insertion (each blood compartment n = 23) or angiographic intervention after TIPS (each blood compartment n = 36) and its relationship with patient outcome was investigated. Results: Angiogenin levels in the inferior caval vein were significantly higher compared to the portal vein (P = 0.048). Ten to 14 days after TIPS, inferior caval vein angiogenin level correlated inversely with the portal systemic pressure gradient (P<0.001), measured invasively during control angiography. Moreover, patients with TIPS revision during this angiography, showed significantly lower angiogenin level in the inferior caval vein compared to patients without TIPS dysfunction (P = 0.01). Conclusion: In cirrhosis patients with complications of severe portal hypertension, circulating levels of angiogenin are derived from the injured liver. Moreover, angiogenin levels in the inferior caval vein after TIPS may predict TIPS dysfunction.
Introduction: Clinically complex patients often require multiple medications. Polypharmacy is associated with inappropriate prescriptions, which may lead to negative outcomes. Few effective tools are available to help physicians optimise patient medication. This study assesses whether an electronic medication management support system (eMMa) reduces hospitalisation and mortality and improves prescription quality/safety in patients with polypharmacy. Methods and analysis: Planned design: pragmatic, parallel cluster-randomised controlled trial; general practices as randomisation unit; patients as analysis unit. As practice recruitment was poor, we included additional data to our primary endpoint analysis for practices and quarters from October 2017 to March 2021. Since randomisation was performed in waves, final study design corresponds to a stepped-wedge design with open cohort and step-length of one quarter. Scope: general practices, Westphalia-Lippe (Germany), caring for BARMER health fund-covered patients. Population: patients (≥18 years) with polypharmacy (≥5 prescriptions). Sample size: initially, 32 patients from each of 539 practices were required for each study arm (17 200 patients/arm), but only 688 practices were randomised after 2 years of recruitment. Design change ensures that 80% power is nonetheless achieved. Intervention: complex intervention eMMa. Follow-up: at least five quarters/cluster (practice). recruitment: practices recruited/randomised at different times; after follow-up, control group practices may access eMMa. Outcomes: primary endpoint is all-cause mortality and hospitalisation; secondary endpoints are number of potentially inappropriate medications, cause-specific hospitalisation preceded by high-risk prescribing and medication underuse. Statistical analysis: primary and secondary outcomes are measured quarterly at patient level. A generalised linear mixed-effect model and repeated patient measurements are used to consider patient clusters within practices. Time and intervention group are considered fixed factors; variation between practices and patients is fitted as random effects. Intention-to-treat principle is used to analyse primary and key secondary endpoints.
DNA methylation-based prediction of response to immune checkpoint inhibition in metastatic melanoma

Background: Therapies based on targeting immune checkpoints have revolutionized the treatment of metastatic melanoma in recent years. Still, biomarkers predicting long-term therapy responses are lacking. Methods: A novel approach of reference-free deconvolution of large-scale DNA methylation data enabled us to develop a machine learning classifier based on CpG sites, specific for latent methylation components (LMC), that allowed for patient allocation to prognostic clusters. DNA methylation data were processed using reference-free analyses (MeDeCom) and reference-based computational tumor deconvolution (MethylCIBERSORT, LUMP). Results: We provide evidence that DNA methylation signatures of tumor tissue from cutaneous metastases are predictive for therapy response to immune checkpoint inhibition in patients with stage IV metastatic melanoma. Conclusions: These results demonstrate that LMC-based segregation of large-scale DNA methylation data is a promising tool for classifier development and treatment response estimation in cancer patients under targeted immunotherapy.
The mammalian target of rapamycin and the integrated stress response are central cellular hubs regulating translation upon stress. The precise proteins and pathway specificity of translation targets of these pathways remained largely unclear. We recently described a new method for quantitative translation proteomics and found that both pathways control translation of the same sets of proteins.
Minimal residual disease (MRD) is the strongest predictor of relapse in B-cell precursor acute lymphoblastic leukemia (BCP-ALL). In BLAST study (NCT01207388), adults with BCP-ALL in remission with MRD after chemotherapy received blinatumomab, a CD19 BiTE® immuno-oncotherapy, 15 µg/m2/day for up to four 6-week cycles (4 weeks continuous infusion, 2 weeks off). Survival was evaluated for 110 patients, including 74 who received HSCT in continuous complete remission. With a median follow-up of 59·8 months, median survival (months) was 36·5 (95% CI: 22.0–not reached [NR]). Median survival was NR (29.5–NR) for complete MRD responders (n = 84) and 14.4 (3.8–32.3) for MRD non-responders (n = 23; p = 0.002); after blinatumomab and HSCT, median survival was NR (25.7–NR) (n = 61) and 16.5 (1.1–NR) (n = 10; p = 0.065), respectively. This final analysis suggests complete MRD response during blinatumomab treatment is curative. Post-hoc analysis of study data suggests while post blinatumomab HSCT may be beneficial in appropriate patients, long-term survival without HSCT is also possible.
Introduction: Dravet syndrome (DS), a prototypic developmental and genetic epileptic encephalopathy (DEE), is characterized by an early onset of treatment-refractory seizures, together with impairments in motor control, behavior, and cognition. Even with multiple conventional anti-epileptic drugs, seizures remain poorly controlled, and there has been a considerable unmet need for effective and tolerable treatments. Areas covered: This targeted literature review aims to highlight recent changes to the therapeutic landscape for DS by summarizing the most up-to-date, evidence-based research, including pivotal data from the clinical development of stiripentol, cannabidiol, and fenfluramine, which are important milestones for DS treatment, together with the latest findings of other pharmacotherapies in development. In phase III, double-blind, placebo-controlled randomized controlled trials stiripentol, cannabidiol, and fenfluramine have shown clinically relevant reductions in convulsive seizure frequency, and are generally well tolerated. Stiripentol was associated with responder rates (greater than 50% reduction in convulsive seizure frequency) of 67%-71%, when added to valproic acid and clobazam; cannabidiol was associated with responder rates of 43%-49% (48%-63% in conjunction with clobazam), and fenfluramine of 54%-68% across studies. Therapies in development include soticlestat, ataluren, verapamil, and clemizole, with strategies to treat the underlying cause of DS, including gene therapy and antisense oligonucleotides beginning to emerge from preclinical studies. Expert opinion: Despite the challenges of drug development in rare diseases, this is an exciting time for the treatment of DS, with the promise of new efficacious and well-tolerated therapies, which may pave the way for treatment advances in other DEEs.
In the age of mechanical reproducibility, the 'aura' surrounding works of art undergoes a crisis. The contemporary relevance of Walter Benjamin's thesis - in its societal, aesthetic, and media-theoretical significance - is illustrated by former U.S. president Donald Trump's purported ownership of "Les deux soeurs" ("Two Sisters"), also known as "Sur la terrasse" ("On the Terrace"), by Pierre-Auguste Renoir. Journalist Mark Bowden, who caught a glimpse of the painting when he was invited to Trump's jet in 1997, describes the event in an article for Vanity Fair: "He showed off the gilded interior of his plane - calling me over to inspect a Renoir on its walls, beckoning me to lean in closely to see … what? The luminosity of the brush strokes? The masterly use of color? No. The signature. 'Worth $10 million,' he told me." Of course, this is the attitude that one might expect from a real-estate mogul turned art collector (although not from a future president). In ignorance of both form and content - to say nothing of their unity - the painting is reduced to its sheer exchange value, concentrated in the signature guaranteeing its authenticity.
In a time when 'internationalization' and 'diversity' have become key areas universities are expected to excel in, it may seem an almost self-evident endeavor to install a memorial for a figure as influential and internationalist as Du Bois, whose connection to the Humboldt University outlasted two ideologically very different political systems. Planned to be positioned in the ground floor of the main building, the memorial, which will start production as soon as the last funding has been secured, reveals an image right at its center that "exist[s] in virtually every student's life and family album, and commonly serve[s] as vehicle[s] of recognition, remembrance and commemoration": the class photograph. What are the main considerations underlying the W. E. B. Du Bois Memorial's concept and design? How has it evolved so far? And what can such a memorial realistically achieve?
The resistance of aesthetics consists in the mode of experience that art affords, which promotes individual consciousness and political awareness by exploding the dualisms with which we tend to simplify things: centralization and decentralization, totality and fragmentation, communism and neoliberal capitalism, dictatorship and democracy. Although the formal complexity and ambiguous compositions met in works by the likes of Picasso, Woolf, and Schönberg most obviously support this sort of experience, it can be drawn out of all art to various degrees. Indeed, what distinguishes these modernists from the artists who came before and after them is how they set aesthetic experience as the aim of artistic production. But no work of art can be reduced either to the whole or to the sum of its parts; either to systematicity or to formlessness. Strictly speaking, the opposing ideals of classical and critical aesthetics are not two distinct aesthetic positions, but the theoretical limits between which art unfolds. By analogy, totalitarian governance and social atomism are not oppositional political materializations, but the two extremes at which politics ends.
The influence and power of some OECD states is under threat but China appears to remain astonishingly flexible, economically potent, and politically strong. How accurate is this view? To answer this question, major aspects of Chinese economic regulation that were adopted in the country’s progress towards capitalist modernization are examined. The analysis requires a historical reconstruction of how China changed the way it intervenes economically and politically, especially with regard to the institutions of the central state. Such a reconstruction reveals that, since the 1990s, the central state has indeed increased its steering capacities. These capacities have a distinctive basis that includes acceptance of a state-centered approach, idiosyncratic innovation policies taking place in the "shadow" of the state’s hierarchy, and the ongoing influence of the communist party. An all-embracing controlling power is, however, not detectable. What does exist in China’s competition-driven system of “statecapitalist” regulation, is a set of limits on the state’s capacity to govern.

Young trees of deciduous Quercus robur and Q. petraea and evergreen Q. ilex were grown together in a competition lysimeter experiment to assess i) – whether the observed growth differences between evergreen Q. ilex and the deciduous Q. robur and Q. petraea on sandy soil in the field and ii) – whether the different natural distribution of Q. robur and Q. petraea could be attributed to physiological differences between the species under experimental drought stress (DS). Half of the plants were subjected to long-term DS in two consecutive years and monitored for physiological and growth parameters. In the first year, water withholding for more than three months did not lead to significant drought stress, probably because of a sufficient residual water volume in the lysimeter for the relatively small plants. However, in the second year, 2018, which was warmer, the bigger plants now competed for the residual water and clear drought stress symptoms developed for more than two months in all trees in the DS lysimeter basin. Growth was only moderately (and mostly not significantly) affected by the DS in the second year, except for a smaller total leaf area in DS Q. ilex as compared to DS Q. robur and Q. petraea and smaller root collar diameter in DS Q. ilex compared to DS Q. robur. Under DS, the deciduous species revealed significant decreases in ΔVIP, indicating a negative effect on electron transport through PS I. Pn, PIabs and water relations parameters (ΨPD and LWC) all decreased to various extents under DS in all three species, leading to clear separation of the deciduous from the evergreen species by PCA. However, PCA did not separate the two deciduous species from each other. It is concluded that longer root growth in the two deciduous species as compared to Q. ilex ameliorates DS effects in Q. robur and Q. petraea and may be the key to understand the better performance of deciduous oaks on sandy soil in the field.
Background: Misconceptions about ADHD stigmatize affected people, reduce credibility of providers, and prevent/delay treatment. To challenge misconceptions, we curated findings with strong evidence base. Methods: We reviewed studies with more than 2000 participants or meta-analyses from five or more studies or 2000 or more participants. We excluded meta-analyses that did not assess publication bias, except for meta-analyses of prevalence. For network meta-analyses we required comparison adjusted funnel plots. We excluded treatment studies with waiting-list or treatment as usual controls. From this literature, we extracted evidence-based assertions about the disorder. Results: We generated 208 empirically supported statements about ADHD. The status of the included statements as empirically supported is approved by 80 authors from 27 countries and 6 continents. The contents of the manuscript are endorsed by 366 people who have read this document and agree with its contents. Conclusions: Many findings in ADHD are supported by meta-analysis. These allow for firm statements about the nature, course, outcome causes, and treatments for disorders that are useful for reducing misconceptions and stigma.
Background: Guselkumab is an interleukin (IL)-23 pathway blocker with proven efficacy in patients with moderate-to-severe plaque psoriasis. Early intervention with guselkumab may result in changes to the clinical disease course versus later intervention. Methods: and analysis Here we present the rationale and design of a phase 3b, randomised, double-blind, multicentre study (GUIDE), comparing treatment effects of guselkumab in patients with short (≤2 years) or longer (>2 years) duration of plaque-type psoriasis, measured from first appearance of psoriatic plaques. Participants achieving skin clearance (Psoriasis Area and Severity Index (PASI)=0) by week 20 and maintaining complete clearance at week 28 visit (‘super-responders’ (SRe)) will be randomised to continue approved maintenance dosing every 8 weeks (q8w) versus an investigational maintenance dosing interval of 16 weeks (q16w) until week 68. Primary endpoint: proportion of participants in the q8w vs q16w arms with absolute PASI <3 at week 68. Participants with PASI <3 at week 68 will be withdrawn from guselkumab treatment for up to 48 weeks. Participants not achieving SRe criteria (non-SRe) will remain in the study with q8w guselkumab dosing through week 68. Additional to serum samples obtained from all patients, skin biopsies and whole-blood samples will be taken from SRe and non-SRe participants at various time points in optional substudies. Analyses include: genetics; immunophenotyping (fluorescence-activated cell sorting); gene and protein expression profiling; immunohistology. By merging clinical endpoints with mechanistic findings, this study aims to elucidate how IL-23 blockade with guselkumab can modify the disease course by altering molecular and cellular drivers that cause relapse after treatment withdrawal, particularly among SRe. Ethics and dissemination: Approval obtained from ethics committee Medical Council Hamburg, Germany (PVN5925). GUIDE is compliant with the Declaration of Helsinki. Trial registration number: Registered at ClinicalTrials.gov (NCT03818035). All primary endpoint results (prespecified analyses) will be submitted to peer-reviewed, international journals within 18 months after primary completion date.
We present the first holographic simulations of non-equilibrium steady state formation in strongly coupled N=4 SYM theory in 3+1 dimensions. We initially join together two thermal baths at different temperatures and chemical potentials and compare the subsequent evolution of the combined system to analytic solutions of the corresponding Riemann problem and to numeric solutions of ideal and viscous hydrodynamics. The time evolution of the energy density that we obtain holographically is consistent with the combination of a shock and a rarefaction wave: A shock wave moves towards the cold bath, and a smooth broadening wave towards the hot bath. Between the two waves emerges a steady state with constant temperature and flow velocity, both of which are accurately described by a shock+rarefaction wave solution of the Riemann problem. In the steady state region, a smooth crossover develops between two regions of different charge density. This is reminiscent of a contact discontinuity in the Riemann problem. We also obtain results for the entanglement entropy of regions crossed by shock and rarefaction waves and find both of them to closely follow the evolution of the energy density.
The Southern Ocean (SO) continental shelf and deep sea are environments characterised by different benthic communities. Their structure and composition are driven and shaped by different variables: whilst on the continental shelf physical environmental variables are the main drivers shaping faunal abundance, structure and composition, the deep-sea fauna is most problably driven by biological variables such as predation and competition. Among shelf and deep-sea benthic communities, peracarids (e.g. amphipods and isopods) are one of the most dominant groups, showing high levels of abundance and diversity in both environments. Knowledge on their assemblage structure and composition in the SO remains limited, as well as the knowledge of the environmental variables that influence them. Therefore, the aim of our study was to investigate peracarid assemblages from the SO continental shelf and deep sea and to assess the main drivers shaping their assemblage structure along a wide bathymetric gradient (from 160 m to about 6000 m depth) and at a large geographic scale. We analysed the spatial distribution of 183,606 peracarids sampled using an epibenthic sledge (EBS) during nine different expeditions in the SO, covering a latitudinal range of 77° to 41° South. Depth was identified as the main driver shaping peracarid abundance pattern, their assemblage structure from the continental shelf (<1499 m) was dissimilar to that from the deep sea (>1500 m). Also, depth was differently correlated with different peracarid orders: while isopod abundances increased with depth, amphipods and mysids were negatively correlated; no correlation was found with cumaceans and tanaidaceans. The dissimilar peracarid assemblage structure between the SO continental shelf and the SO deep sea can be due to the assumption that there are different driving forces shaping benthic assemblages from these two environments (physical variables on the continental shelf, biological interactions in the deep sea). As a result, we also suggest that environmental changes due to climate change (e.g. temperature, ice coverage, productivity) would have different consequences depending on the bathymetric range considered.
Circulating P2X7 receptor signaling components as diagnostic biomarkers for temporal lobe epilepsy

Circulating molecules have potential as biomarkers to support the diagnosis of epilepsy and to assist with differential diagnosis, for example, in conditions resembling epilepsy, such as in psychogenic non-epileptic seizures (PNES). The P2X7 receptor (P2X7R) is an important regulator of inflammation and mounting evidence supports its activation in the brain during epilepsy. Whether the P2X7R or P2X7R-dependent signaling molecules can be used as biomarkers of epilepsy has not been reported. P2X7R levels were analyzed by quantitative ELISA using plasma samples from controls and patients with temporal lobe epilepsy (TLE) or PNES. Moreover, blood cell P2X7R expression and P2X7R-dependent cytokine signature was measured following status epilepticus in P2X7R-EGFP reporter, wildtype, and P2X7R-knockout mice. P2X7R plasma levels were higher in TLE patients when compared with controls and patients with PNES. Plasma levels of the broad inflammatory marker protein C-Reactive protein (CRP) were similar between the three groups. Using P2X7R-EGFP reporter mice, we identified monocytes as the main blood cell type expressing P2X7R after experimentally evoked seizures. Finally, cytokine array analysis in P2X7R-deficient mice identified KC/GRO as a potential P2X7R-dependent plasma biomarker following status epilepticus and during epilepsy. Our data suggest that P2X7R signaling components may be a promising subclass of circulating biomarkers to support the diagnosis of epilepsy.
In 1887, Mayr described four species of the ant genus Strumigenys collected in the Brazilian state of Santa Catarina: Strumigenys unidentata, Strumigenys subedentata, Strumigenys denticulata, and Strumigenys crassicornis. All of them were described based on a series of one to several specimens, without designation of a holotype, as usual at that time. The same can be said about Strumigenys eggersi, described by Emery in 1890 based on specimens collected in Saint Thomas (U.S. Virgin Islands), without designation of a holotype. In 1961, Brown designated a lectotype for S. unidentata and synonymized it under Strumigenys louisianae. However, the specimens belonging to the type series of the other four species remain as syntypes. Considering that these are four of the most frequently collected species of Strumigenys in the Neotropical region, in this work we provide lectotype designations and complete redescriptions for S. crassicornis, S. denticulata, S. eggersi, and S. subedentata to ensure the taxonomic stability of these names.
Chemosensory impairments have been established as a specific indicator of COVID-19. They affect most patients and may persist long past the resolution of respiratory symptoms, representing an unprecedented medical challenge. Since the SARS-CoV-2 pandemic started, we now know much more about smell, taste, and chemesthesis loss associated with COVID-19. However, the temporal dynamics and characteristics of recovery are still unknown. Here, capitalizing on data from the Global Consortium for Chemosensory Research (GCCR) crowdsourced survey, we assessed chemosensory abilities after the resolution of respiratory symptoms in participants diagnosed with COVID-19 during the first wave of the pandemic in Italy. This analysis led to the identification of two patterns of chemosensory recovery, partial and substantial, which were found to be associated with differential age, degrees of chemosensory loss, and regional patterns. Uncovering the self-reported phenomenology of recovery from smell, taste, and chemesthetic disorders is the first, yet essential step, to provide healthcare professionals with the tools to take purposeful and targeted action to address chemosensory disorders and their severe discomfort.
Objective: The NADPH oxidase Nox4 is an important source of H2O2. Nox4-derived H2O2 limits vascular inflammation and promotes smooth muscle differentiation. On this basis, the role of Nox4 for restenosis development was determined in the mouse carotid artery injury model. Methods and results: Genetic deletion of Nox4 by a tamoxifen-activated Cre-Lox-system did not impact on neointima formation in the carotid artery wire injury model. To understand this unexpected finding, time-resolved single-cell RNA-sequencing (scRNAseq) from injured carotid arteries of control mice and massive-analysis-of-cDNA-ends (MACE)-RNAseq from the neointima harvested by laser capture microdissection of control and Nox4 knockout mice was performed. This revealed that resting smooth muscle cells (SMCs) and fibroblasts exhibit high Nox4 expression, but that the proliferating de-differentiated SMCs, which give rise to the neointima, have low Nox4 expression. In line with this, the first weeks after injury, gene expression was unchanged between the carotid artery neointimas of control and Nox4 knockout mice. Conclusion: Upon vascular injury, Nox4 expression is transiently lost in the cells which comprise the neointima. NADPH oxidase 4 therefore does not interfere with restenosis development after wire-induced vascular injury.
FAD synthase is the last enzyme in the pathway that converts riboflavin into FAD. In Saccharomyces cerevisiae, the gene encoding for FAD synthase is FAD1, from which a sole protein product (Fad1p) is expected to be generated. In this work, we showed that a natural Fad1p exists in yeast mitochondria and that, in its recombinant form, the protein is able, per se, to both enter mitochondria and to be destined to cytosol. Thus, we propose that FAD1 generates two echoforms—that is, two identical proteins addressed to different subcellular compartments. To shed light on the mechanism underlying the subcellular destination of Fad1p, the 3′ region of FAD1 mRNA was analyzed by 3′RACE experiments, which revealed the existence of (at least) two FAD1 transcripts with different 3′UTRs, the short one being 128 bp and the long one being 759 bp. Bioinformatic analysis on these 3′UTRs allowed us to predict the existence of a cis-acting mitochondrial localization motif, present in both the transcripts and, presumably, involved in protein targeting based on the 3′UTR context. Here, we propose that the long FAD1 transcript might be responsible for the generation of mitochondrial Fad1p echoform.
Glutathione has long been suspected to be the primary low molecular weight compound present in all cells promoting the oxidative protein folding, but twenty years ago it was found “not guilty”. Now, new surprising evidence repeats its request to be the “smoking gun” which reopens the criminal trial revealing the crucial involvement of this tripeptide.
We analyze the ESG rating criteria used by prominent agencies and show that there is a lack of a commonality in the definition of ESG (i) characteristics, (ii) attributes and (iii) standards in defining E, S and G components. We provide evidence that heterogeneity in rating criteria can lead agencies to have opposite opinions on the same evaluated companies and that agreement across those providers is substantially low. Those alternative definitions of ESG also affect sustainable investments leading to the identification of different investment universes and consequently to the creation of different benchmarks. This implies that in the asset management industry it is extremely difficult to measure the ability of a fund manager if financial performances are strongly conditioned by the chosen ESG benchmark. Finally, we find that the disagreement in the scores provided by the rating agencies disperses the effect of preferences of ESG investors on asset prices, to the point that even when there is agreement, it has no impact on financial performances.
The sum of Lyapunov exponents Lf of a semi-stable fibration is the ratio of the degree of the Hodge bundle by the Euler characteristic of the base. This ratio is bounded from above by the Arakelov inequality. Sheng-Li Tan showed that for fiber genus g≥2 the Arakelov equality is never attained. We investigate whether there are sequences of fibrations approaching asymptotically the Arakelov bound. The answer turns out to be no, if the fibration is smooth, or non-hyperelliptic, or has a small base genus. Moreover, we construct examples of semi-stable fibrations showing that Teichmüller curves are not attaining the maximal possible value of Lf.
Background: To identify variables predicting outcome in neuroblastoma patients assigned to the high-risk group solely by the presence of MYCN oncogene amplification (MNA). Methods: Clinical characteristics, genomic information, and outcome of 190 patients solely assigned to high-risk neuroblastoma by MNA were analyzed and compared to 205 patients with stage 4 neuroblastoma aged ≥18 months with MNA (control group). Results: Event-free survival (EFS) and overall survival (OS) at 10 years were 47% (95%-CI 39–54%) and 56% (95%-CI 49–63%), respectively, which was significantly better than EFS and OS of the control group (EFS 25%, 95%-CI 18–31%, p < 0.001; OS 32% 95%-CI 25–39%, p < 0.001). The presence of RAS-/p53-pathway gene alterations was associated with impaired 10-year EFS and OS (19% vs. 55%, and 19% vs. 67%, respectively; both p < 0.001). In time-dependent multivariable analyses, alterations of RAS-/p53-pathway genes and the extent of the best primary tumor resection were the only independent prognostic variables for OS (p < 0.001 and p = 0.011, respectively). Conclusions: Neuroblastoma patients attributed to high risk solely by MYCN amplification have generally a more favorable outcome. Mutations of genes of the RAS and/or p53 pathways and incomplete resection are the main risk factors predicting poor outcome.
Background: Bipolar disorder is associated with circadian disruption and a high risk of suicidal behavior. In a previous exploratory study of patients with bipolar I disorder, we found that a history of suicide attempts was associated with differences between winter and summer levels of solar insolation. The purpose of this study was to confirm this finding using international data from 42% more collection sites and 25% more countries. Methods: Data analyzed were from 71 prior and new collection sites in 40 countries at a wide range of latitudes. The analysis included 4876 patients with bipolar I disorder, 45% more data than previously analyzed. Of the patients, 1496 (30.7%) had a history of suicide attempt. Solar insolation data, the amount of the sun’s electromagnetic energy striking the surface of the earth, was obtained for each onset location (479 locations in 64 countries). Results: This analysis confirmed the results of the exploratory study with the same best model and slightly better statistical significance. There was a significant inverse association between a history of suicide attempts and the ratio of mean winter insolation to mean summer insolation (mean winter insolation/mean summer insolation). This ratio is largest near the equator which has little change in solar insolation over the year, and smallest near the poles where the winter insolation is very small compared to the summer insolation. Other variables in the model associated with an increased risk of suicide attempts were a history of alcohol or substance abuse, female gender, and younger birth cohort. The winter/summer insolation ratio was also replaced with the ratio of minimum mean monthly insolation to the maximum mean monthly insolation to accommodate insolation patterns in the tropics, and nearly identical results were found. All estimated coefficients were significant at p < 0.01. Conclusion: A large change in solar insolation, both between winter and summer and between the minimum and maximum monthly values, may increase the risk of suicide attempts in bipolar I disorder. With frequent circadian rhythm dysfunction and suicidal behavior in bipolar disorder, greater understanding of the optimal roles of daylight and electric lighting in circadian entrainment is needed.

Where Haas sees the narrative dividing into "Streberwitz" and "Kriegsdarstellung" I see something more like a division between 'Witz' and 'Krieg' per se. The point and the provocation of the novel, in my view, is that Kehlmann declines to bring these two strata together, or rather: that he first insists on bringing them together, by forcing Tyll and the Thirty Years War to inhabit the same work, and then refuses to synthesize them into anything like a higher unity. The irony of the fool, in Tyll, does not acquire gravity or depth by virtue of its relationship to a reality whose hidden truths it emphatically does not reveal; and the reality of war does not find redemption or sublimation in art.
Like identical twins, philosophy and history seem to be tied together in an uneasy way. On the one hand, philosophy is very concerned to engage with the history of philosophy. There are not many other branches of knowledge so preoccupied with continually referring back to their own 'classics'. On the other hand, quite a few of these classical authors did not hold history in high esteem. Aristotle, as is well known, even preferred drama to history, arguing that the latter merely concerned contingent issues. The marriage between history and philosophy quite often results in monsters like Hegelian philosophy of history: grand narratives that are all too easy to criticize and to debunk. If we want to better understand this complex relationship between philosophy and history, it might be worth turning to the German philosopher Hans Blumenberg.
The formation of terrestrial planets was a complex process which begun in the very early stage of the Solar System in the protoplanetary disk (PPD). Chondrites are fragments of planet precursors, which have never experienced differentiation and can help to reconstruct the first processes leading to planet formation. The main components of chondrites are chondrules, calcium-aluminum-rich inclusions (CAIs), amoeboid olivine aggregates (AOAs), metals and fine-grained material. Each of these components formed by a complex mechanism involving aggregation and/or melting. Previous research has already provided an overall view of the formation of these objects, however, there are still open questions regarding the aggregation behavior of particles, the heating mechanism(s) and the thermal history of CAIs, AOAs and chondrules. For instance, the involvement of flash-heating events and electrostatics in the aggregation and melting of these objects has been a keen topic of discussion.
The aim of this doctoral thesis was to develop and carry out an experiment to study various early Solar System processes under long-term microgravity. In the project with the acronym EXCISS (Experimental Chondrule Formation aboard the ISS), free-floating, 126(23)µm-sized Mg2SiO4 dust particles were exposed to electric fields and electric discharges.
The experimental set-up was installed inside a 10x10x15 cm3-sized container and consisted of an arc generation unit connected to the sample chamber, a camera with an optical system, a power supply unit with lithium-ion batteries and the EXCISS mainboard with a Raspberry Pi Zero and mass storage devices. The sample chamber was manufactured from quartz glass and the experiments were filmed. The complete experiment container was subsequently returned to the Goethe University and the samples were analyzed with scanning electron microscopy, electron backscatter diffraction and synchrotron micro-CT.
Video analysis has shown that particles, which were agitated by electric discharges, align in chains within the electric field with their longest axis parallel to the electric field lines. Consequently, electric fields could have influenced the inner structure and porosity of particle aggregates in the PPD.
The discharge experiments produced fused aggregates and individual melt spherules.
The fused aggregates share many morphological characteristics with natural fluffy-type CAIs and some igneous CAIs found in chondrites. Consequently, CAIs could have formed by the aggregation of particles with various degrees of melting. Further, a small amount of melting could have supplied the required stability for such fractal structures to have survived transportation and aggregation to, and subsequent compaction within, developing planetesimals.
Some initial particles were completely melted by the arc discharges and formed melt spherules. The newly formed olivines crystallized with a preferred orientation of the [010] axis perpendicular to the surface of the spherule. Similar preferred orientations have been found in natural chondrules. However, the microstructure differs from the results of previous experiments on Earth, which show, for example, crystal settling on one side of the sample because of the influence of gravity. Furthermore, the melt spherules show evidence for an interaction of the melt with the surrounding hot gas. Therefore, microgravity experiments with more advanced experimental parameters bear great potential for future chondrule formation experiments.
If one thing can be learned from the recent boom in the apparently 'new' field of the 'history of the humanities', it is that, especially in the humanities, the history of an academic discipline is never mere history, because the research questions that inaugurate a discipline continue to subsist at its foundations. Knowledge in the humanities, it seems, develops differently. In many fields, 'progress' is far less linear than in the natural sciences; indeed, research programmes may shuttle back and forth between different epochs, with interpretations of the past continually shedding new light upon the present.
In recent years, several neuronal differentiation protocols were published that circumvent the requirement of embryoid body (EB) formation under serum-deprivation and simplified medium conditions. But a neuronal default model to establish an approach that works efficiently for all pluripotent cells and neuronal precursors is still lacking. Whether such a default neural mechanism exist and how this is implemented across a broad spectrum of cell source, is addressed in several studies and still controversially discussed. It was proposed that the default neuronal fate is initiated in the absence of extrinsic signals and is achieved by eliminating extracellular inhibitors of neuroectodermal fate and suppressing cell-cell signalling through limited cell density. Previous studies reported that ESC and ECC grown at low density and in absence of exogenous factors or feeder layers die within 24 h but acquire a neural identity as indicated by expression of the neural marker Nestin. Thus, this application is not suitable for generating neural cultures. Furthermore, it was reported that P19 cells survive and express neuroectodermal marker genes in serum-free DMEM/F12 medium containing transferrin, insulin, and selenite, although no neurites were identified.
Based on this background, in this study, a novel approach to induce neuronal differentiation in vitro was developed that implements a nutrient-poor environment, which, in contrast to previous studies, ensures the survival of neuronally differentiated cells over a long period of time and allows normal formation of neurites. Neither the formation of free-floating aggregates nor supplementation of growth factors or known inducers was required to establish a reliable neuronal differentiation protocol. A simple medium, consisting of DMEM/F12+N2 that was highly diluted in salt solution, was sufficient to drive a fast neuronal differentiation in monolayer cultures. Serum deprivation and strong dilution of DMEM/F12+N2 medium cause a nutrient-poor environment in which the influence of growth factors and inducers is minimized. This medium creates a metabolically defined environment that is presumably free of extrinsic signals that prevent the decision of neuronal fate. Analysis of the medium components discovered no actual inducer. Hence, it was suggested that the metabolic composition of the medium exclusively covers specific cell requirements of neurons, therefore ensures their survival, and drives the switch from pluripotent cells to neurons. The self-developed method was established by usage of the murine embryonal carcinoma cell line P19 and could be transferred to murine ESC. Consequently, the method could provide a feasible protocol for a generally valid neuronal default model.
The established protocol provides several advantages such as the possibility to generate stable pure neuronal cultures by a fast, simple, and highly reproducible one-step induction under defined medium conditions with a minimum of exogen effectors. The method is characterised by clear and steady medium conditions that makes the investigation of specific cell requirements during differentiation accessible. It is therefore expected to be a useful tool to investigate the molecular basis of neuronal differentiation as well as for high throughput screenings. The phenotype of mature postmitotic neurons was arising within one week and cultures were shown to stay stable at least for three weeks. The neuronal identity was confirmed by expression of neuronal markers through immunofluorescence staining and mass spectrometry analysis. Furthermore, increased levels of axon markers were detected in early neuronal differentiation and functionality of the synapses of the P19-derived neurons was ascertained by detection of calcium activity. Axonal laser ablation, immediately followed by fast regrowth of connections in the neuronal network, revealed a strong regeneration potential under the given conditions. Furthermore, the generated neurons showed a morphologically distinct phenotype and the formation of neural rosettes. Immunofluorescence staining demonstrated the generation of pure and homogeneous neuronal cultures, free of glial cells.
Retinoic acid (RA) plays an essential role in cell signalling during embryogenesis and efficiently induces neuronal differentiation in vitro in a concentration dependent manner. Neither retinol nor retinoic acid was included in any of the components of the self-prepared medium in this work. However, I observed, dependence on RARβ- and/or RARγ-regulated RA signalling in serum-free monolayer cultures. Nevertheless, neuronal differentiation in serum-free monolayer cultures was assumed to be RARα-independent because (i) RARα was slightly downregulated after neuronal induction, (ii) the truncated RARα of the RAC65 mutant had no effect on induction efficiency, and (iii) a pan-RAR inhibitor suppressed neuronal differentiation. In contrast to serum-free monolayer cultures, the truncated RARα prevented neuronal differentiation by application of the conventional protocol where cells are grown in free floating cell aggregates in serum-containing medium. Proteome analysis of P19 cells, treated by the self-developed differentiation protocol over five days showed increased levels of cellular RA binding proteins that mediate the cellular RA transport and are involved in canonical as well as non-canonical RA signalling.

The conception of the whole as a system, that is, as a totality determined by one principle or idea, has dominated the philosophical tradition from Kant and Hegel to Marxism - and, as Louis Althusser's critique of Hegelianism shows, not without implicit social, political, and ideological consequences. The possibility of breaking with the idealist tradition in all of these respects rests on the articulation of an alternative conception of the whole. Althusser advances the notion of the social whole as a complex unity that is constituted through its own effects - what he calls "overdetermination." Such overdetermination of the whole displaces the conception of the whole as totality (Hegel) in favor of Spinoza's notion of modal unity - the whole as singularity.
Diverse museum diversities

'Diversity' has become a lively key word in contemporary museum discourse and practice, with numerous policies and initiatives being conducted under its banner. Achieving 'diversity' is seen as something to be celebrated - a good thing in itself. But quite what 'diversity' refers to is itself heterogeneous, with this only rarely explicitly articulated or even recognised. As such, what exists is a shifting field of diverse diversities, which variously interlink and reinforce each other but which may also mask critical discrepancies, disconnects, incompatibilities and even contrary ambitions.
The book deals with a comprehensive constellation of narrative and visual, often counterposed representations of the causes, course, and results of the assault on the Palace of Justice of Colombia by a guerrilla commando and the immediate counterattack launched by state security forces on November 6, 1985, as well as with the local memorial traditions in which the production, circulation and reproduction of these representations have taken place between 1985 and 2020. The research on which it is based was grounded in the method and perspective of classical anthropology, in as much as qualitative fieldwork and the search for the perspective of the actors involved have played a central role. Within that context, memory entrepreneurs belonging to diverse sectors, from the far-right to the human rights movement, were followed through multisited fieldwork in various locations of Colombia, as well as in various countries of America and Europe. The analyses of fieldwork data, documental sources, and visual representations that constitute the core of the argument are framed in the field of memory studies and mainly based on theoretical and methodological resources from Pierre Bourdieu’s Field Theory, Jeffrey Alexander’s theory of social trauma, and Ernst Gombrich’s characterization of iconological analysis.
The book is composed of four chapters preceded by an introduction and followed by the conclusions and documental appendices, and substantiates three main theses. The first is that the Palace of Justice events were a radio- and television-broadcasted dispersed tragedy that affected the lives of actors from different social sectors and regions of Colombia, who have launched since 1985 multiple memorial initiatives in different fields of culture, thereby contributing to the formation and intergenerational transmission of a widespread cultural trauma. The second is that the narrative and visual representations at the core of that trauma express a vast universe of local representational traditions that can be traced at least until the early 20th century, and therefore preexists the so-called Colombian “memory boom”, dated to the mid-1990s. As an example of the preexistence and longstanding impact of these traditions, the local usage of the figure of “holocaust” for representing the effects of politically motivated violence is analyzed regarding the Palace of Justice events, but also traced to other representations emerged in the decade of 1920. The third thesis is that analyzing the diverse, frequently counterposed accounts of political violence elaborated within these traditions provides an opportunity to explore a wide variety of understandings of the causes and characteristics of the longstanding Colombian social and armed conflict.

Introduction: The newest intravenous (IV) iron products show an improved safety profile over predecessors, allowing for the rapid administration of relatively high doses. Ferric derisomaltose (FDI; also known as iron isomaltoside), ferric carboxymaltose (FCM), and ferumoxytol (FER), are successful treatments for iron deficiency (Europe; FDI and FCM) and iron deficiency anemia (US; FDI, FCM, and FER). Areas covered: This review focusses on the chemistry and structure of FDI, FCM, and FER, and on three key aspects of IV iron safety: (1) hypersensitivity; (2) hypophosphatemia and sequelae; (3) cardiovascular safety. Expert opinion: Although the safety of modern IV iron has improved, immediate infusion reactions and the development of hypophosphatemia must be appreciated and recognized by those who prescribe and administer IV iron. Immediate infusion reactions can occur with any IV iron and are usually mild; severe reactions – particularly anaphylaxis – are extremely rare. The recognition and appropriate management of infusion reactions is an important consideration to the successful administration of IV iron. Severe, persistent, hypophosphatemia is a specific side effect of FCM. No cardiovascular safety signal has been identified for IV iron. Ongoing trials in heart failure will provide additional long-term efficacy and safety data.
