Die kumulative Dissertation beschäftigt sich mit der atmosphärischen Konzentration von Eiskeimen, einer Unterklasse des atmosphärischen Aerosols, die bei der Eisbildung in Wolken eine zentrale Bedeutung besitzt. Messungen der Eiskeimkonzentration am Taunusobservatorium (Kleiner Feldberg) (nahe Frankfurt am Main) wurden mit dem Verfahren einer Vakuum-Diffusionskammer durchgeführt. Die Arbeit umfasst die Darstellung des angewandten Messverfahrens und die Analyse und Bewertung der Messergebnisse für den Raum Zentraleuropa, anhand von u.a. Rückwärtstrajektorien und Korrelationen zu aerosolphysikalischen Parametern. Ein signifikanter Einfluss von Mineralstaub-Ferntransport aus Wüstengebieten auf die Eiskeimkonzentration in Zentral-Europa wurde ermittelt.
This dissertation consists of three essays, which study the relation between stock prices and the macroeconomy using vector autoregressions (VARs). The first essay focuses on the link between stock prices and the current account. I find that stock markets provide a channel, in addition to the traditional exchange rate channel, through which external balance for a country with a current account imbalance can be restored. The second essay explores the transmission of U.S. stock price shocks to real activity and prices in G-7 countries. I achieve identification by imposing a small number of sign restrictions on impulse responses, while controlling for monetary policy, business cycle and government spending shocks. The results suggest that stock price movements are important for fluctuations in G-7 real activity and prices, but do not qualify as demand side business cycle shocks. The third essay investigates the impact of monetary and technology shocks on the stock market. I find an important role for technology shocks, but not monetary shocks, in explaining variations in real stock prices. The identification method is flexible enough to study the effects of technology news shocks. The responses are consistent with the idea that news on technology improvements have an immediate impact on stock prices.
Das Chronic Care Model (CCM) stellt eine Zusammenfassung evidenzbasierter Erkenntnisse dar, um die Versorgung chronisch Erkrankter zu optimieren. Diverse Messinstrumente sind in den letzten Jahren zur Erfassung des Implementierungserfolges des CCM aus Sicht der Leistungserbringer und der Patienten entwickelt worden. Einen Ansatz zur Erfassung, inwiefern die im CCM beinhalteten Dimensionen angewendet werden, stellt das Patient Assessment of Chronic Illness Care (PACIC- 5A) dar. Dieses Instrument wurde im englischsprachigen Raum entwickelt und primär im amerikanischen Gesundheitssystem eingesetzt. Es besteht in der ursprünglichen Fassung aus 20 Items und erfragt in fünf Subskalen CCM-konkordante Aspekte: Patientenaktivierung, Leistungserbringerstruktur / Entscheidungshilfen, Zielsetzung, Problemlösung / Beratung, Follow-up / Koordination. Die erweiterte Version aus 26 Items erlaubt zusätzlich zur Beurteilung der Erfüllung des CCM die Analyse der Umsetzung im Sinne des 5A-Modells, eines behaviouristischen Ansatzes zur Verhaltensänderung. Ziel dieser Arbeit war es, den Fragebogen in die deutsche Sprache ohne Verlust des semantischen Inhaltes zu übersetzen und an einer primärmedizinischen Population im deutschen Gesundheitssystem zu evaluieren. Eine weitere Zielsetzung war, den Implementierungsgrad des CCM-Konstruktes im spezifischen Fall der Major Depression zu erkunden. Hierzu wurde das übersetzte PACIC-5A im Rahmen der im Institut für Allgemeinmedizin der Universitätsklinik Frankfurt durchgeführten ProMPT-Studie zur Betreuung von Patienten mit Major Depression an insgesamt 509 Patienten erprobt. Hiervon wurden 436 Patienten in die Studie eingeschlossen. Hierauf erfolgte die Überprüfung der psychometrischen Eigenschaften. Die statistischen Messergebnisse, hinsichtlich innerer Konsistenz (Cronbach´s α) und Trennschärfe (korrigierte Item-Skala-Korrelation), ergaben gute und mit der aktuellen Literatur übereinstimmende Resultate. Die konvergente Validität des PACIC-5A wurde anhand des primärmedizinisch validen Fragebogens EUROPEP (European Project on Patient Evaluation of General Practice Care) geprüft. Das Ergebnis zeigte hohe Korrelationen vor allem in den inhaltlich vergleichbaren Skalen, wie z.B. Skala „Patientenaktivierung“ aus dem PACIC-5A und Skala „Information und Unterstützung“ des EUROPEP. Die diskriminierenden Fähigkeiten des Fragebogens wurden durch die geringen korrelativen Eigenschaften mit Skalen des EUROPEP, welche andere Dimensionen erörtern, nachgewiesen. Zusätzlich ermöglichte der Vergleich der Mittelwerte der einzelnen Skalen des PACIC-5A zwischen dem Kontroll- und Interventionsarm eine zusätzliche Beurteilung der diskriminierenden Potenz. Das PACIC-5A ist bis jetzt vorrangig an Patienten mit chronischen Erkrankungen aus dem organischen Formenkreis (z.B. Osteoarthritis, Diabetes Mellitus) angewendet und validiert worden. Die kongruente Anwendung des PACIC-5A auf Erkrankte mit Major Depression ist ein limitierender Aspekt. Hier besteht weiterer Forschungsbedarf. Für eine optimale Patientenversorgung in der Primärmedizin ist es essentiell, primärmedizinisch valide Instrumente den Hausärzten zur Verfügung zu stellen.
Aim: To study the changes in leiomyoma volume following uterine artery embolization (UAE) and to correlate these changes with the initial leiomyoma volume and location within the uterus and to evaluate the impact of preprocedural prediction of the best tube angle obliquity for visualization of the uterine artery origin using 3D-reconstructed contrast-enhanced MR angiography (CE-MRA) on the radiation dose, fluoroscopy time and contrast medium volume used during UAE. Materials and Methods: The study was performed in two parts. The first part was retrospectively done on 28 patients (age range: 37-57 years, mean: 48 years, SD: 4.81) in whom UAE was performed. All leiomyomas in all patients were evaluated. In total, 84 leiomyomas were evaluated. MRI studies were performed before, 3 months and 1 year after UAE. The volumes and location of each leiomyoma in each patient were evaluated in consensus by two radiologists. The second part included 40 consecutive patients (age range: 37-56 years, mean: 46 years, SD: 4.49) and was done in a controlled prospective/retrospective manner. In 20 sample patients (prospective part) pre-procedural prediction of the best tube angle obliquity was predicted using 3D-reconstructed CE-MRA and provided to the interventionalist. 3D-reconstruction was done using Inspace application. The radiation dose, fluoroscopy time and contrast medium volume for those patients were compared with the data of the last 20 procedures (control) performed by the same interventionalist (retrospective part). Results: For the first part the mean pre-embolization volume was 51.6 cm3 range:0.72-371.1cm3, SD=79.3). At 3-month follow-up 83 (98.8%) leiomyomas showed a mean volume reduction of 52.62% (range: 12.79–96.67%, SD=21.85) and 1 leiomyoma (1.2%) increased in volume. At 1-year follow-up 5 (6%) leiomyomas were not detectable, 72 (85.7%) showed a further mean of 20.5% (range: 2.52–58.72%, SD=11.92) volume reduction compared to the 3-month follow-up volume and 7 (8.3%) leiomyomas increased in volume. A statistically significant (p=0.026 at 3-month, p=0.0046 at 1-year) difference in percentage of volume change was observed based on leiomyoma location; submucous leiomyomas showed the largest volume reduction. The initial leiomyoma volume showed a weak negative correlation (Spearman's correlation-coefficient =-0.35 at 3m and -0.36 at 1y) with the leiomyoma volume change. For the second part the tube angle prediction resulted in a significant reduction of the radiation dose utilized (p<0.001), fluoroscopy time (p=0.002) and contrast medium volume (p<0.001) for the sample patients when compared with the control patients. The overall radiation dose was reduced from a mean of 11044 μGym2 to a mean of 4172.5 μGym2, fluoroscopy time was reduced from a mean of 15.45 minutes to 8.81 minutes and contrast medium volume was reduced from a mean of 135 ml to 75 ml. Conclusion: UAE results in significant leiomyoma volume reduction at 3-month and 1- year follow-up. The leiomyoma location plays an important role in volume changes while the initial leiomyoma volume plays a minor role. Pre-procedural prediction of the best tube angle obliquity for visualization of the origin of the uterine artery using 3D-reconstructed CE-MRA results in a significant reduction of the radiation dose, fluoroscopy time and contrast medium volume used during UAE.
Although throughout the history of anthropology the ethnography of urban societies was never an important topic, investigations on cities in Africa contributed to the early theoretical development of urban studies in social sciences. As the ethnography of rural migrants in towns made clear, cultural diversity and creativity are foundational and permanent elements of urban cultures in Africa (and beyond). Currently, two new aspects complement these insights: 1) Different forms of mobility have received a new awareness through the concept of transnationalism. They are much more complex, including not only rural–urban migration, but also urban–urban migration, and migrations with a destination beyond the continent. 2) Urban life-worlds also include the appropriation of globally circulating images and lifestyles, which contribute substantially to the current cultural dynamics of cities in Africa. These two aspects are the reasons for the high complexity of urban contexts in Africa. Therefore, whether it is still appropriate to speak about the “locality” of these life-worlds has become questionable. At the same time, these new aspects explain the self-consciousness of members of urban cultures in Africa. They contribute to the expansive character of these societies and to the impression that cities in Africa host the most innovative and creative societies worldwide.
With which political developments is BiKF confronted as a research centre as well as concerning its research and transfer efforts? Are there any hints for emerging research questions that meet practical needs? This paper gives an overview – as of June 2010 – on priority issues in the run-up to CBD’s COP-10, the 10th Conference of the Parties to the Convention on Biological Diversity (CBD), which will take place in Nagoya/Japan in October 2010. Highlighted discourse threads are: (1) the state of negotiations for an Access and Benefit Sharing (ABS) regime within CBD, (2) European and international preparations for renewing the political objectives for protecting biodiversity (Post-2010 Targets) and (3) the recent decision on an Intergovernmental Science-Policy Platform for Biodiversity and Ecosystem Services (IPBES). These three threads are selected against the background of an in depth analysis of the discourse field which was carried out in 2008/09 for BiKF. They show how the field progresses and which developments are worth being incorporated into BiKF’s further work. This Knowledge Flow Paper documents the talk given by the author during the second BiKF Retreat, 17–18 July 2010.
Die Ausbreitung von HIV stellt ein kontinuierlich wachsendes Problem dar [132]. Durch Einführung der hochaktiven antiretroviralen Therapie (HAART) konnte die Morbidität und Mortalität der HIV-Infektion deutlich gesenkt werden, jedoch limitieren Resistenzbildungen des Virus und Toxizität der Medikamente den Erfolg. Eine mögliche Therapiealternative bietet die HIV-Gentherapie. Hierbei werden Zellen eines Patienten genetisch modifiziert, so dass sie ein antivirales Genprodukt exprimieren. In der Arbeitsgruppe von Laer (Georg-Speyer-Haus, Frankfurt) wurde der retrovirale Vektor M87o entwickelt, der das antivirale, membranverankerte Peptid maC46 kodiert. Dieses hemmt als Fusionsinhibitor effizient den Viruseintritt von HIV. Als Zielzellen einer HIV-Gentherapie können neben TLymphozyten, den eigentlichen Zielzellen von HIV, auch deren Vorläufer, die hämatopoetischen Stammzellen, verwendet werden. Durch Generierung der gesamten Hämatopoese sollte dies zur Expression des antiviralen Transgens in allen Blutzelllinien führen. Besonders wichtig hierbei ist, dass die Funktion der hämatopoetischen Stammzellen durch die genetische Modifikation möglichst nicht gestört wird. Ziel der vorliegenden Arbeit war es daher, toxische Effekte von M87o auf die Repopulierungsfähigkeit hämatopoetischer Stammzellen auszuschließen. Neben den Toxizitätsanalysen sollte auch die Langzeitexpression des retroviralen Vektors nach Transplantation genetisch modifizierter T- und Stammzellen untersucht werden. Eine stabile Expression des Transgens ist vor allem in T-Lymphozyten als Hauptzielzellen von HIV ausschlaggebend für den Erfolg der Gentherapie. Daher war ein weiteres Ziel dieser Arbeit, die Transgenexpression in vivo besonders in T-Lymphozyten im Verlauf zu untersuchen. Hierzu wurden in einem syngenen Mausmodell hämatopoetische Stammzellen mit dem retroviralen Vektor M87o transduziert und in bestrahlte Rag1-defiziente Mäuse transplantiert. Damit mögliche toxische Effekte von M87o auf die Hämatopoese nicht durch den Anteil untransduzierter Zellen im Transplantat maskiert werden, wurde in einer Versuchgruppe der Anteil transduzierter Stammzellen durch MACS-Sortierung auf über 95% angehoben. bAls Kontrollgruppen wurden untransduzierte, aber gleichermaßen kultivierte Stammzellen sowie mit dem Kontrollvektor M87c transduzierte Stammzellen transplantiert. Im folgenden Beobachtungszeitraum von 18-20 Wochen wurde regelmäßig das periphere Blut der Empfängertiere analysiert sowie nach Tötung der Tiere die einzelnen Zellpopulationen der hämatopoetischen Organe Blut, Lymphknoten und Milz charakterisiert. Hierbei konnte keine Toxizität durch M87o nachgewiesen werden. Zwar wurde für M87o-angereicherte Stammzelltransplantate eine verminderte bzw. verzögerte Lymphozytenrepopulierung beobachtet, dies war jedoch wahrscheinlich auf eine eingeschränkte „Fitness“ der Stammzellen durch den Sortierungsprozess und eine geringere Zellzahl im Transplantat zurückzuführen. M87o-transduzierte Stammzellen waren schließlich in der Lage, die komplette Lymphopoese zu generieren. Im Blut, Lymphknoten und Milz der Rezipienten konnten NK-, T- und B-Zellen nachgewiesen werden. Die lymphatische Differenzierung wurde also durch M87o nicht beeinträchtigt. Eine Aussage über die Toxizität von M87o auf die Myelopoese konnte leider nicht getroffen werden. Nach subletaler Bestrahlung der Empfängertiere und damit nur teilweisen Ablation des endogenen Knochenmarks wurden die meisten Zellen der myeloischen Linie durch die Wirts-Stammzellen generiert. Es müssen somit hinsichtlich der Unbedenklichkeit von M87o noch weitere präklinische Untersuchungen erfolgen, bei denen durch letale Bestrahlung der Empfängertiere lediglich die durch Spenderzellen differenzierte Myelopoese analysiert werden kann. Bei den Untersuchungen zur Transgenexpression nach Transplantation genetisch modifizierter Stammzellen konnte eine Langzeitexpression des maC46-Peptids auf allen lymphatischen Zelllinien (T-, B- und NK-Zellen) nachgewiesen werden. Dies zeigt also, dass eine stabile und effiziente Integration des Transgens und somit eine langfristige Expression in vivo möglich ist. Im Verlauf konnten jedoch bei nahezu allen Tieren fallende Anteile M87o-exprimierender Lymphozyten nachgewiesen werden. Dieser beobachtete Expressionsverlust war variabel hinsichtlich des zeitlichen Auftretens sowie zelltypabhängig. Die höchsten Anteile M87o-exprimierender Zellen zeigten sich innerhalb der B-Lymphozyten. Im Rahmen der M87o-Expressionsanalyse nach Transplantation genetisch modifizierter T-Lymphozyten wurden T-Lymphozyten mit unterschiedlicher Transduktionseffizienz in Rag1-defiziente Mäuse transplantiert. Unterschiede in der Langzeitexpression in Abhängigkeit von der ins Genom integrierten Kopienzahl des Vektors konnten hierbei nicht eindeutig gezeigt werden. Bei einigen Tieren konnte eine relativ langfristige in vivo Expression des maC46-Peptids nachgewiesen werden, bei anderen hingegen nachlassende Transgenexpressionen. Insgesamt war die Aussagekraft hier jedoch durch eine nach Transplantation auftretende schwere Kolitis bei den Versuchstieren und somit limitierte Beobachtungszeit stark eingeschränkt.
Das West-Nil-Virus (WNV) gehört zur Gruppe des Japan-Enzephalitis-Serokomplexes der Flaviviren, welches in Afrika, Asien und Mittleren Osten sowie in Süd- und Osteuropa seine Verbreitung findet. Das unerwartete Auftreten des Erregers in den USA (New York, 1999) bedingte eine große Zahl an transfusions- sowie transplantations-assoziierten WNV-Infektionen. Derzeit liegen keine Erkenntnisse über die Verbreitung des WNV in der deutschen Blutspenderpopulation vor. Aus diesem Grund war das Ziel dieser Arbeit die Bestimmung der Prävalenz und Inzidenz des Erregers zur Einschätzung der Gefahr von transfusions-assoziierten WNV-Infektionen. Zunächst wurde zur Bestimmung der Prävalenz des Erregers die Sensitivität und Spezifität verschiedener ELISAs (Panbio, Focus und Euroimmun) für die WNV-Antikörpertestung bestimmt. Es zeigte sich, dass WNV starke Kreuzreaktionen mit dem für unsere Breiten relevanten Frühsommer-Meningoenzephalitis-Virus (FSME) aufweist. Aus diesem Grund wurde zusätzlich ein FSME-ELISA sowie als Bestätigungstest ein Neutralisationstest verwendet und ein Testalgorithmus entwickelt. Für die Bestimmung der Inzidenz wurde auf den Grundlagen von Hadfield eine WNV-Reverse Transkriptase-PCR (RT-PCR) entwickelt und etabliert. Die 95% Nachweisgrenze wurde nach der Entwicklung der RT-PCR auf 54Kop./ml WNV-RNA bezogen auf eine Einzelprobe in einem Minipool bestehend aus acht Blutspenden bestimmt. Das Konfidenzintervall beträgt (CI): 42; 6 - 79; 92 Kopien/ml. Zudem wurde die Sensitivität der automatischen Extraktion mittels des auf magnetischen Festphasen beruhenden Separationsmoduls I mit der WNV-RT-PCR ermittelt. Die 95% Nachweisgrenze lag bei 225Kop./ml WNVRNA((CI): 168; 5 - 314; 3 ) bezogen auf eine Einzelprobe in einem Minipool bestehend aus acht Blutspenden. Aufgrund der etwas schlechteren Sensitivität wurde für die Bestimmung der Inzidenz die manuelle Extraktion durchgeführt. Zudem wurde das kommerzielle WNV Procleix Assay der Firma Chiron verwendet, um alle bisher in Europa aufgetretenen WNV-Stämme zu erfassen. Im Sommer 2004 wurde zunächst die Prävalenz des WNV bestimmt. Hierfür wurden 14:000 aus Hessen stammende Blutspenden von gesunden Spendern untersucht. Initial zeigte sich eine hohe Rate an anti-WNV reaktiven (5; 9 %) Blutspenden, wobei nur 0; 03% der Blutspenden im Neutralisationstest als Anti-WNV positiv bestätigt worden. 0; 15% der bestätigten anti-WNV positiven Blutspenden waren hierbei am ehesten mit Reisen in WNV-Epidemiegebiete assoziiert. Die Bestimmung der Inzidenz erfolgt im darauf folgendem Jahr durch die Untersuchung von 10:976 Blutspenden zusammengefasst in 1:372 Minipools mittels WNV RT-PCR. Von 1:372 Minipools wurden 1:247 Minipools mit dem Procleix WNV Assay untersucht. Beide Untersuchungsmethoden konnten keine WNV-positive Blutspende nachweisen. Insgesamt scheint das WNV -wenn überhaupt- nur in verschwindend geringem Maße in der deutschen Blutspenderpopulation zu exisitieren. Somit besteht derzeit keine Gefahr für transfusions-assoziierte WNV-Infektionen. Dennoch wurden importierte WNV-Infektionen aus Endemiegebieten in Deutschland beschrieben. Durch weitere Veränderungen der klimatischen Gegebenheiten wäre die Einschleppung des WNV nach Deutschland in Zukunft durchaus denkbar. Hierfür steht nun eine für das Spenderscreening taugliche Real-time PCR zür Verfügung, so dass jederzeit ein Blutspenderscreening auf WNV eingeführt werden kann.
Somsanit® (Natrium-γ-Hydroxybutyrat) wird schon seit vielen Jahren zur Langzeitsedierung von Intensivpatienten eingesetzt. Durch das Vorliegen als Natriumsalz, kann bei Langzeitanwendung oft schon nach wenigen Tagen eine Hyperatriämie auftreten, die zum Absetzen des Präparates führen kann. Um diesen Nachteil von Somsanit® zu überwinden, wurde von Dr. F. Köhler Chemie das Natrium-freie GHB-Analogon Butamidol (γ-Hydroxybuttersäure-Ethanolamid) entwickelt. In klinischen Studien zeigte Butamidol eine adäquate Sedierung von Intensivpatienten, besaß aber selbst in hohen Dosierungen keine narkotische Wirkung. GHB, welches auch physiologisch in Gehirn vorkommt, führt konzentrationsabhängig von einer tiefen Sedation bis hin zur Narkose. Aus der Literatur ist bekannt, dass beide GHB-Effekte durch einen Agonismus an GABAB-Rezeptoren ausgelöst werden. Die GHB-Konzentrationen die dafür benötigt werden, können aber nur durch exogen zugeführtes GHB erreicht werden. In physiologischen Konzentrationen bindet GHB an den GHB-Rezeptor, dessen Funktion bisher noch nicht eindeutig aufgeklärt werden konnte. Das Ziel dieser Arbeit war, die Affinität von Butamidol und GHB an den beiden Systemen GABAB- und GHB-Rezeptor zu untersuchen und mögliche Unterschiede aufzuklären. Im Radiorezeptor-Assay zeigte Butamidol eine geringere Affinität zum GABAB-Rezeptor als GHB (Butamidol: Ki = 399.5 mM; GHB: Ki = 10.6 mM) und eine agonistische Wirkung am GABAB-Rezeptor wurde, über die Hemmung spannungsabhängiger Ca2+-Kanäle in Synaptosomen, nur für GHB nachgewiesen. In der nachfolgenden Untersuchung der GABAB-Rezeptor Signaltransduktionskaskade wurden, durch die akute GHB-Gabe, die Phospho-ERK-Level im Hippocampus von Mäusen vermindert. Der gleiche Effekt wurde für die akute Behandlung mit dem GABAB-Agonisten Baclofen erhalten, aber nicht für Butamidol. In Experimenten zur Modulation des GABAergen Systems, durch die subchronische Behandlung von Mäusen mit GHB oder Butamidol, wurde nur nach GHB-Behandlung eine Steigerung des synaptosomalen GABA-Uptakes detektiert. Somit konnte eine Beeinflussung des GABAergen Systems durch Butamidol an mehreren möglichen Angriffspunkten ausgeschlossen werden. Als zweites in Frage kommendes Target für Butamidol wurde das GHB-System untersucht. In Radiorezeptor-Bindungs-Versuchen wurden zur Verdrängung des antagonistischen Radioliganden [3H]NCS-382 höhere Butamidol Konzentrationen benötigt, als zur Verdrängung des Agonisten [3H]GHB (Ki: 1296 vs. 452 μM). Ein vergleichbarer „Shift“ zu höheren Konzentrationen zeigte auch der „kalte“ Agonist GHB und nicht der „kalte“ Antagonist NCS-382 (GHB: Ki 3.10 vs. 1.41 μM; NCS-382: Ki 0.431 vs. 0.447 μM). In einer weiteren subchronischen Behandlungsstudie von Ratten mit Butamidol und GHB wurde für GHB eine Erniedrigung der Gesamt-Rezeptor-Population und für Butamidol eine Erhöhung des Anteils der Agonist-Bindungsstellen an der Gesamt-Rezeptor-Population erhalten. Dieses Ergebnis kann für GHB als Hinweis auf ein agonistisches und für Butamidol als partiell agonistisches Bindungsverhalten am GHB-Rezeptor interpretiert werden. Da im GHB-Radio-Rezeptor-Assay relativ hohe Butamidol Konzentrationen zur Verdrängung der Radioliganden eingesetzt werden mussten, wurden daran anschließend Untersuchungen zur klinischen Relevanz, der für die GHB-Rezeptor Bindung notwendigen Butamidol Konzentrationen, durchgeführt. In Behandlungsstudien von Mäusen, mit anschließender Bestimmung der Butamidol Plasma- und Hirnspiegel, wurde die Hirn/Plasma Ratio ermittelt. Aus der erhaltenen Hirn/Plasma Ratio und den aus klinischen Studien bekannten beim Menschen wirksamen Plasma-Spiegeln, lassen sich für Butamidol Hirnkonzentrationen von 200-350 μM extrapolieren, die im Konzentrationsbereich für die Bindung von Butamidol am GHB-Rezeptor liegen.
In der Therapie des Diabetes mellitus Typ 2 ist neben Patientenschulung, Fewichtsreduktion und körperlicher Aktivität die Therapie mit oralen Antidiabetika von besonderer Bedeutung, um langfristig die Blutzuckereinstellung zu verbessern und kardiovaskuläre wie andere Folgeerkrankungen zu minimieren. Allerdings sinkt im Verlauf die Effizienz dieser Therapie, so dass die dauerhafte Gabe von Insulin notwendig wird. Dabei können jedoch auch schwerwiegende Nebenwirkungen, wie z.B. Hypoglykämien und eine wiederrum mit einem erhöhten Risiko an kardiovaskulären Erkrankungen verbundene ausgeprägte Gewichtszunahme auftreten. Demgegenüber führte der GLP-1 Rezeptor-Agonist Exenatide in jüngsten Studien zu einer Verbesserung der Blutzuckereinstellung und Reduktion des Körpergewichts und kann somit als eine neuartige Therapiealternative zum bisher verwendeten Insulin angesehen werden. In der hier vorliegenden prospektiven Studie wurde Exenatide mit Insulin bei Typ 2-Diabetes Patienten mit einer unzureichenden Stoffwechseleinstellung unter Metformin in Bezug auf deren Blutzuckereinstellung und das Hypoglykämie-Risiko verglichen. In dieser 26-wöchigen, multizentrischen, kontrollierten, randomisierten und zweiarmigen Phase IIIb-Studie wurden insgesamt 494 Patienten aus ganz Deutschland untersucht. Die Patienten mit einer unzureichenden Metformin- (Primärkollektiv) oder Kombinationstherapie (exploratives Kollektiv: Metformin plus Sulfonylharnstoff) wurden im Verhältnis 3:1 auf zweimal täglich Exenatide oder zweimal täglich Mischinsulin Aspart 30/70 randomisiert. In einem hierarchischen Modell wurde zunächst die Nichtunterlegenheit von Exenatide gegenüber dem Mischinsulin im Hinblick auf das primäre Endziel der Blutzuckereinstellung (in Form des HbA1c-Wertes) untersucht und anschließend die Überlegenheit von Exenatide in Hinblick auf ein geringeres Hypoglykämie-Risiko. Sekundäre Studienziele waren die Häufigkeit von schweren und von nächtlichen Hypoglykämien, die Veränderung des Körpergewichts sowie des Body Mass Index. Ferner wurden 7-Punkt-Blutzuckertagesprofile und Patientenfragebögen in die Auswertung einbezogen. Bei der insgesamt in acht Visiten unterteilten Studie erfolgte nach zwei Wochen die Randomisierung auf die beiden Therapie-Arme. Während die Dosierung des Mischinsulins im Verlauf der gesamten Studiendauer vom Arzt individuell titriert werden konnte, wurde die Dosis von Exenatide nach vier Wochen von 2 x 5 μg/Tag auf 2 x 10 μg/Tag verdoppelt und bis zum Ende der Studie nicht verändert. Die hier vorliegende Arbeit ist die erste Studie, die das Hypoglykämie-Risiko unter Exenatide und Mischinsulin als primären Endpunkt prospektiv und konfirmatorisch vergleichend untersucht. Im Primärkollektiv konnte die Nicht-Unterlegenheit von Exenatide zum Mischinsulin in Bezug auf den HbA1c-Wert festgestellt werden. Bei der Vermeidung von Hypoglykämien war Exenatide dem Mischinsulin statistisch signifikant überlegen. Zusätzlich konnte unter der Exenatide-Behandlung ein signifikanter Gewichtsverlust festgestellt werden, wohingegen die Patienten im Mischinsulin-Arm signifikant an Gewicht zunahmen. Die gleichzeitige Erreichung dieser Therapieziele (HbA1c<7,0%, keine Gewichtszunahme und keine Hypoglykämien) wurde als Triple-Endpoint definiert und im Exenatide-Arm signifikant häufiger als unter der Mischinsulin-Therapie beobachtet. In den 7-Punkt-Blutzuckertagesprofilen konnte neben einer allgemeinen Blutzuckersenkung in beiden Therapiearmen eine zusätzliche postprandiale Senkung zu den Zeiten der morgendlichen und abendlichen Exenatide-Injektion beobachtet werden. Demgegenüber waren die präprandialen Blutzuckerwerte im Mischinsulin-Arm niedriger, so dass insgesamt eine vergleichbare Verbesserung des Flächenintegrals in den Blutzuckertageskurven erzielt wurde, was die vergleichbare HbA1c-Verbesserung erklärt. Entsprechend zum Gewicht nahmen auch BMI und Hüftumfang der Patienten unter Exenatide ab und unter Insulin zu. Das Gesamtcholesterin und die Triglyceride sanken unter beiden Therapien leicht ab, HDL-Cholesterin stieg leicht an, wohingegen nur unter Exenatide das LDL-Cholesterin abnahm. Ebenfalls sanken nur im Exenatide-Arm der systolische und diastolische Blutdruck. Die häufigsten unerwünschten Ereignisse unter Exenatide waren Übelkeit, Erbrechen, Verdauungsstörungen, Durchfall, Kopfschmerzen und Erkältungen, wobei Durchfall, Kopfschmerzen und Erkältungen auch unter Insulin zu beobachten waren. Bei der in den Patientenfragebögen DTSQ und SF-12 dokumentierten subjektiven Empfindung der Nebenwirkungen zeigten sich allerdings eine Kompensation der deutlicheren Nebenwirkungen von Exenatide durch dessen Vorteile, u.a. im Hinblick auf seine einfachere Dosierung und der als angenehm empfundenen Gewichtsreduktion. Insgesamt war in beiden Therapie-Armen eine ausreichende Patientenzufriedenheit festzustellen. Unter denjenigen Patienten, bei denen Antikörper gegen Exenatide festgestellt wurden, nahm die Prävalenz einer Antikörperbildung gegen Exenatide bis zum Ende der Studie ab. Die Ergebnisse der hier vorgelegten Studie lassen die Schlussforderung zu, dass der GLP-1 Agonist Exenatide eine therapeutische Alternative zum Mischinsulin in der Behandlung des Diabetes Mellitus Typ 2 darstellt. Da eine Restfunktion der pankreatischen ß-Zellen eine Voraussetzung für den Therapieerfolg darstellt, sollte Exenatide möglichst früh bei nachlassender Wirkung des oralen Antidiabetikums Metformin eingesetzt werden. Hier wirkt Exenatide durch eine Verbesserung der Blutzuckereinstellung, die zudem mit einem verminderten Hypoglykämie-Risiko und einer Gewichtsreduktion verbunden ist. Die Kombination dieser drei bedeutenden Effekte hat den klinisch relevanten Vorteil, das Risiko an kardiovaskulären Spätfolgen und somit die Morbidität und Mortalität von Typ 2-Diabetikern langfristig zu senken. Dieses sollte in einer prospektiven Langzeitstudie untermauert werden.
Das Ziel der vorliegenden Arbeit war der Einbau, die Inbetriebnahme, die Abstimmung und der Test eines Strahlmatchingsystems in eine Zweistrahl-RFQ-Beschleunigerstruktur. Dieses Strahlmatchingsystem wurde entwickelt, um die Beschleunigereinheit des Frankfurter Funneling-Experimentes besser an die nachfolgende HF-Deflektoreinheit anzupassen und um zu zeigen, dass ein Strahlmatching innerhalb der RFQ-Beschleunigerstruktur möglich ist. Des Weiteren wurden die zum Versuch gehörigen Ionenquellen modifiziert, um eine bessere Anpassung der Strahlherstellung an die Beschleunigerstruktur zu erreichen. Die Spannungsverteilung in der Beschleunigerstruktur selbst wurde durch weitergehende Tuningmaßnahmen verbessert, um die Teilchenverluste weiter zu minimieren Mit dem Funnelingexperiment soll experimentell geprüft werden, ob eine Strahlstromerhöhung durch das Zusammenführen mehrerer Ionenstrahlen verschiedener Ionenquellen möglich ist. Solch ein System ist für einige Zukunftsprojekte (HIDIF, SNS-Ausbau, ESS, u.ä.), die große Strahlströme benötigen, die nicht aus nur einer Ionenquelle extrahiert werden können, erforderlich. Das in dieser Arbeit behandelte Strahlmatching ist für das Experiment notwendig, da zu große Teilchenverluste in der Funnelsektion entstanden und somit eine bessere Anpassung des Strahls an den HF-Deflektor erforderlich wurde. Es konnte gezeigt werden, dass die hier verwendete Art der Strahlfokussierung auch in eine komplizierte RFQ-Beschleunigerstruktur integrierbar ist, in der zwei Strahlkanäle auf der gleichen Stützen-Bodenplatten-Konstruktion aufgebaut sind. Die Verlängerung der Endelektroden und die Integration einer Strahlanpassung haben einen positiven Einfluss auf die Transsmission innerhalb des Beschleunigers und verbessern die Transmission durch den Deflektor. Es konnten Energiemessungen und zeitaufgelöste Faradaytassenmessungen der Teilchenbunche sowie zeitaufgelöst Makropulse mit der Faradaytasse gemessen werden. Floureszensschirmmessungen zeigten, dass die beiden Teilchenstrahlen auf eine neue gemeinsame Strahlachse gebogen wurden. Die Energiemessung zeigte, dass die Simulationen mit RFQSIM sehr genau die Endenergie der Teilchen berechnen konnte. Im Strahlkreuzungspunkt hinter dem Zweistrahl-RFQ-Beschleuniger konnten nahezu identische Teilchenbunche erzeugt werden. Diese Teilchenbunche wiesen zudem die in vorherigen Simulationen errechneten Charakteristika auf, in denen eine transversale und eine longitudinale Fokussierung gegenüber dem ungematchten Strahl simuliert wurden. Es konnte auch eine weitere Strahlradiusreduzierung gemessen werden, die auf eine exaktere Justierung der Elektroden zurückzuführen ist. Die Phasenfokussierung konnte verbessert werden, indem die Elektrodenspannung besser an die Strahlmatchingsektion angepasst wurde. Hierzu mussten auch die Einschussparameter der Strahlen in die Beschleuniger angepasst werden, damit die Transmission der Beschleuniger sich nicht verschlechterte. Insgesamt konnte mit den durchgeführten Experimenten erstmals demonstriert werden, dass zwei Strahlen in einem RFQ-Beschleuniger auf einen Punkt hinter dem Beschleuniger angepasst werden können und die Spannungsverteilung in solch einer Struktur durch Tuningmaßnahmen abstimmbar ist. Es konnte erstmals demonstriert werden das über 90% der Teilchen beider Strahlen, bei guten Strahleigenschaften, auf eine neue gemeinsame Strahlachse abgelenkt (gefunnelt) wurden.
Das Ziel der vorliegenden Arbeit war, zu analysieren, welche Einflussgrößen sich auf die Kryotransferergebnisse positiv oder negativ auswirken können, um somit Paaren, die über kryokonservierte Zellen verfügen, am effektivsten zu einem gemeinsamen Kind zu verhelfen.
Untersucht wurde, ob das Alter der Patientinnen, die Art der Befruchtung, die Anzahl der übertragenen Embryonen und die Zyklusvorbereitung einen Unterschied auf die Entwicklungs- und Implantationsfähigkeit menschlicher Eizellen sowie auf die Schwangerschaftsrate nach Kryokonservierung und Auftauen im Pronucleus-Stadium aufwiesen.
In der vorliegenden Studie wurden 126 Patientinnen mit ungewollter Kinderlosigkeit, die zwischen 2002 und 2004 am Universitätsklinikum Frankfurt am Main in Behandlung waren, aufgenommen. Das Durchschnittsalter der Patientinnen lag bei 33,3 Jahren. Von 226 durchgeführten Kryotransferzyklen resultierten die Pronukleus-Eizellen bei 34 Kryotransferzyklen aus einer In-vitro-Fertilisation (IVF) und bei 192 Zyklen aus einer intrazytoplasmatischen Spermieninjektion (ICSI).
Es wurde festgestellt, dass in der vorliegenden Studie die erzielte Erfolgsrate, gemessen an der relativ geringen Schwangerschaftsrate, deutlich niedriger lag als beim deutschen IVF-Register. Das Resultat muss jedoch auch unter dem Aspekt der schlechten Ergebnisse der IVF-/ICSI-Therapie in der untersuchten Gruppe betrachtet werden.
Die Ergebnisse der Kryotherapie sind dagegen mit den Zahlen anderer Reproduktionszentren vergleichbar, die derzeit eine Effektivität von durchschnittlich 19,12 % erreichen (DIR 2007) (23).
Bei der Untersuchung wurde jeweils nur der erste und zweite Kryotransferzyklus pro Patientin ausgewertet, damit die Voraussetzung zur Untersuchung der Einflüsse anamnestischer Faktoren gleich war.
Durch ICSI befruchtete Eizellen führten nach Kryokonservierung seltener zu einer Schwangerschaft (11,59%) als nach IVF (16,66%), beim Primärtransfer war das Ergebnis weniger verschieden (9,41% und 7,40%).
Die Vorbereitung des Kryotransferzylus war insofern relevant, als das der Transfer im spontanen Zyklus bezüglich der Schwangerschaftsrate am erfolgreichsten war (13,79%). Der Transfer in einem artifiziellen Zyklus, also nach Stimulation aus einer Kombination von HMG mit FSH, der alleinigen Verabreichung beider Medikamente oder nach dem Kaufmann-Schema, erbrachte dagegen nur eine geringe Schwangerschaftsrate von 6,45%.
Signifikante Unterschiede wiesen die im Punktionszyklus verwendeten Medikamente zur ovariellen Stimulation hinsichtlich der Degenerationsrate nach dem Auftauen der PN-Stadien auf, die nach HMG-Stimulation niedriger (19,35%) als nach FSH-Stimulation (30,82%) war, die Schwangerschaftsrate wies allerdings keine deutlichen Unterschiede auf (11,90% und 13,08%).
Das Alter der Patientinnen hatte wider Erwarten in der vorliegenden Untersuchung keinen eindeutigen Einfluss auf die Schwangerschaftsrate.
Die Resultate bestätigten, dass ein Transfer von maximal drei erlaubten Embryonen die besten Ergebnisse erzielte (19,6 %), hingegen bei einem Transfer von zwei Embryonen ein Ergebnis von nur 5,5 % vorlag. Dieses Ergebnis spricht, wie bereits bei anderen Forschungsergebnissen postuliert, für einen Transfer von drei Embryonen und sollte somit angestrebt werden.
Es lässt sich zusammenfassen, dass die Ergebnisse der Kryotransferbehandlungen in Bezug auf die untersuchten Parameter, wie spontan/hormonelle Kryotransferzyklen, IVF- oder ICSI- Behandlung und Alter, nur geringe Unterschiede aufwiesen.
Einen signifikanten Einfluss auf die Erfolgsrate hatten jedoch die Anzahl der transferierten Embryonen und die verschiedenen Stimulationsmedikamente auf die Degeneration der Oozyten.
Die vorliegende Arbeit hat gezeigt, dass die Kryokonservierung die Effizienz der reproduktionsmedizinischen Behandlung nachweislich erhöht. Darüber hinaus plädiert sie für ein Überdenken des deutschen Embryonenschutzgesetzes, um somit den Frauen eine hohe Anzahl von Behandlungszyklen zu ersparen.
Ziel dieser Arbeit war die Aufklärung lichtinduzierter Strukturänderungen verschiedener photoschaltbarer Moleküle durch zeitaufgelöste Infrarotspektroskopie. Hierzu war es notwendig, eine komplexe Messapparatur zu konzipieren, aufzubauen und zu optimieren. Das entwickelte Anreg-/Abtast-Experiment ermöglicht die Messung kleinster transienter Absorptionsänderungen (delta A<1E-5) im Spektralbereich von 1000 cm^-1 bis 2500 cm^-1 mit einer Zeitauflösung von etwa 0,3 ps. Es können Anregungspulse im Bereich von 258nm bis über 600nm generiert werden. Über ein computergesteuertes Wellenplättchen kann die Polarisation des Anregungslichtes während der Datenaufnahme variiert werden, was eine Auswertung hinsichtlich der molekularen Anisotropie ermöglicht. Die eingesetzten Probenzellen gestatten die Untersuchung geringster Probenmengen (V <20µL) bei Temperaturen bis zu 50°C. Nitrophenylacetat (NPA) wurde hinsichtlich seiner Photodecarboxylierungsreaktion untersucht. Für alle drei Konstitutionsisomere konnte nachgewiesen werden, dass die Freisetzung von CO2 innerhalb von 1 ns erfolgt. Es zeigen sich signifikante Unterschiede zwischen den Reaktionsverläufen der drei Konstitutionen, wobei die erhaltenen Amplituden der Photoproduktspektren mit den bekannten Decarboxylierungsquantenausbeuten korrelieren. Die globale Analyse ergibt einen multiexponentiellen Aufbau des CO2-Signals. Im Fall von meta- und para-NPA erfolgt die Abspaltung von CO2 über einen dominanten Zerfallskanal mit einer Zeitkonstante von t~200 ps. Quantenchemische Rechnungen legen nahe, dass dieser Hauptreaktionspfad über den Triplettzustand verläuft. Bei ortho- NPA wird dieser Zerfallskanal effektiv gequencht, was mit einer schnellen Deaktivierung des angeregten Singulettzustandes durch einen intramolekularen Protonentransfer erklärt wird. Nach diesen Messungen steht fest, dass meta-Nitrophenylacetat hervorragend als caged compound für CO2 geeignet ist. Die Primärdynamik von solubilisiertem Proteorhodopsin (PR) in D2O wurde im infraroten und sichtbaren Spektralbereich sowohl bei saurem als auch bei alkalischem pD-Wert untersucht. Dies erlaubt den direkten Vergleich der in beiden Spektralbereichen gemessenen Daten. Der primäre Protonenaktzeptor Asp97 liegt dabei entweder protoniert (pD=6,4) oder deprotoniert (pD=9,2) vor. Die transienten vis-Absorptionsspektren ergaben, wie bei PR in H2O, einen biexponentiellen Zerfall des angeregten Zustands und die gleichzeitige Bildung des PRK-Photoproduktes. Die Abweichung der bei PR in D2O ermittelten Werte von den publizierten Zeitkonstanten der H2O-Messung wird als kinetischer Isoptopeneffekt interpretiert. Dieser variiert mit dem pD-Wert, was auf Unterschiede der Wasserstoffbrückenbindungsnetzwerke in der Retinalumgebung hinweist. Die Signaturen der transienten Infrarotspektren werden den C=C- und C=N-Moden des Retinals sowie der Amid I-Mode des Proteins zugeordnet. Es konnte ebenfalls die Bildung des PRK-Produktes nachgewiesen werden, wobei die Isomerisierungsquantenausbeute des Retinals unabhängig von der Protonierung von Asp97 ist. Die im IR bestimmten Zeitkonstanten sind dabei unabhängig vom pD-Wert und weichen von den im Sichtbaren ermittelten Werten ab. Dieser Befund wird mit dem Einfluss der molekularen Temperatur auf die transienten IR-Spektren und dem Auftreten von Kühlprozessen erklärt. Zusätzlich zum Wildtyp-Protein wurde die PR-D97N-Mutante als Modellsystem für PR im sauren pH-Bereich ebenfalls im vis- und IR-Bereich untersucht. Die dabei erzielten Ergebnisse stehen im Einklang mit den bisherigen Ausführungen. Der geringe kinetische Isotopeneffekt weist auf ein ähnliches Wasserstoffbrückennetzwerk wie beim Wildtyp unter sauren Bedingungen hin. Als letztes wurde ein synthetisches Modellcollagen untersucht. Die Seitenkettenverbrückung der Peptidsequenz mit einem Azobenzol-basierten, künstlichen Photoschalter sollte eine lichtinduzierte Entfaltung der Tertiärstruktur ermöglichen. Der isolierte Photoschalter und die gekoppelte Sequenz wurden zunächst unter Gleichgewichtsbedingungen sowohl im UV/vis- als auch im IR -Bereich umfangreich spektroskopisch charakterisiert. Diese Messdaten lagen zum großen Teil bereits vor und wurden in dieser Arbeit einer detaillierten Auswertung unterzogen. Für den isolierten Schalter konnte im IR-Bereich eine umfassende Bandenzuordnung erstellt werden. Dabei wird im photostationären Gleichgewicht eine reversible trans-> cis-Isomerisierung festgestellt, welche keine Temperaturabhängigkeit aufweist. Darüberhinaus wurden polarisationsabhängige, transiente IR-Spektren des isolierten Azoschalters für die trans->cis- und die cis->trans-Isomerisierungsrichtung aufgenommen. Die instantan auftretenden, markanten Differenzsignale können den Amid I- und Amid II-Moden der im Schalter enthaltenen Peptidbindungen sowie den Phenylmoden zugeordnet werden. Die extrahierten kinetischen Parameter sind für beide Isomere nahezu identisch, was durch einen dominanten Beitrag molekularer Kühlprozesse erklärt werden kann. Aufgrund der geringen Isomerisierungsquantenausbeute (< 10 %) können die Differenzspektren der photostationären Gleichgewichte nicht in den Produktspektren dieser Messungen reproduziert werden. Hinsichtlich der ermittelten Anisotropieparameter ergeben sich kleine Unterschiede zwischen beiden Datensätzen. Zusammen mit theoretischen Modellierungen werden diese in Zukunft genauere Aussagen über die Strukturen der Isomere erlauben. Die UV/vis-Absorptionsspektren des gekoppelten Systems zeigen, dass die Absorptionsbanden des Azoschalters durch die Kopplung an das Collagen nicht signifikant beeinflusst werden. Im IR-Absorptionsspektrum konnten wichtige Amid I-, Amid II- und Amid II0-Banden des Azoschalters und der Peptidsequenz sowie eine große Anzahl weiterer Banden zugeordnet werden. Temperaturabhängige Absolut- und Differenzspektren im UV/vis- und IR-Bereich zeigen eine irreversible thermische Denaturierung der Collagentripelhelix ab etwa 50°C. Das verwendete, deuterierte Lösungsmittelgemisch führt zu einem H/D-Austausch. Anhand der Amid II-Bande der in der tripelhelikalen Struktur geschützten Glycine kann die Existenz der Collagenstruktur und ihre Entfaltung nachgewiesen werden. Die Amplituden der photostationären Differenzspektren sind jedoch kleiner als beim isolierten Schalter, was auf eine verringerte Isomerisierungsquantenausbeute hinweist. Die bei Erwärmung beobachtete Vergrößerung der Differenzsignale wird mit einer effizienteren Isomerisierung nach dem Aufschmelzen der Tripelhelix erklärt. Für die trans->cis-Isomerisierungsrichtung wurden transiente IR-Spektren des Azocollagens bei unterschiedlichen Temperaturen aufgenommen. Alle instantan auftretenden Differenzsignale können auf die Schwingungsmoden des Azoschalters zurückgeführt werden. Bei einer Temperatur von 50°C lässt sich ein Einfluss der Peptidsequenz auf die transienten Spektren nicht nachweisen, was mit einer aufgeschmolzenen Tertiärstruktur im Einklang ist. Hingegen wird bei 20°C das Ausbleichen von Amid I-Schwingungsbanden der Peptidsequenz beobachtet, was eindeutig einen Energietransfer auf diese Moden zeigt. Bei 35°C sind die Bleichsignale des Collagens in diesem Bereich bereits deutlich abgeschwächt. Die transienten Spektren des isolierten Azoschalters besitzen keine derartige Temperaturabhängigkeit.
Orthopockenviren sind große DNA-Viren, die im Zytoplasma der Wirtszelle replizieren und für über 200 Proteine kodieren. Sie besitzen ein breites Wirtszellspektrum (host-range) und modulieren auf komplexe Art und Weise zelluläre Prozesse, um ihre Replikation zu gewährleisten. Zu diesem Genus der Familie der Pockenviren gehört auch das modifizierte Vacciniavirus Ankara (MVA). MVA ist ein hoch attenuiertes, replikationsdefizientes Impfvirus, dem im Vergleich zu ursprünglichen Vacciniavirus-Stämmen viele virale Genfunktionen fehlen. Zu diesen verlorengegangenen Genen zählen so genannte host-range-Gene, die für das breite Wirtszellspektrum des Vacciniavirus (VACV) verantwortlich sind, deren molekulare Funktion aber größtenteils unbekannt ist. Diese Arbeit befasste sich zum einen mit der Untersuchung der Rolle der host-range-Gene K1L und C7L in der MVA-Infektion. Zum anderen sollte geprüft werden, ob der im MVA-Genom unvollständige Leserahmen F11L durch Wiederherstellung seiner Funktionalität den Wirtstropismus von MVA erweitern kann. Das Fehlen von K1L und C7L in MVA ist mit dem Verlust der späten viralen Genexpression verbunden. Als mögliche Ursache hierfür wurde in dieser Arbeit die Phosphorylierung des eukaryotischen Translationsinitiationsfaktors 2alpha (eIF2alpha) entdeckt, welche zum Abbruch der Proteinsynthese in der infizierten Zelle führt. Unter den möglichen Kinasen wurde die Proteinkinase R (PKR) als das verantwortliche Schlüsselenzym identifiziert und somit gezeigt, dass das K1- und C7-Protein den anti-viralen PKR-eIF2alpha-Signalweg inhibieren. Es stellte sich heraus, dass die eIF2alpha-Phosphorylierung alleine jedoch nicht für das Fehlen der späten Genexpression verantwortlich ist. Neben dem inhibitorischen Einfluss auf den PKR-eIF2alpha-Signalweg zeigte sich, dass C7 die Aktivierung des NFKB-Signalwegs reduziert, welcher für eine anti-virale Antwort der Wirtszelle wichtig ist. Ein weiterer Ansatz zur Aufklärung der K1- und C7-Funktion bestand darin, zelluläre Interaktionspartner zu identifizieren. Hierbei konnte das heterogene nukleäre Ribonukleoprotein K (HNRPK) als möglicher Interaktionspartner von C7 entdeckt werden. Neben den bisher bekannten host-range-Genen gibt es vermutlich weitere Gene, die den Wirtsbereich des VACV definieren. Das im MVA-Genom defekte F11L-Gen war ein guter Kandidat für eine solche Genfunktion, da es bei der Virionenmorphogenese, Virusausbreitung und der Migration VACV-infizierter Zellen eine Rolle zu spielen scheint. Deshalb wurde ein rekombinantes MVA mit vollständiger F11L-Gensequenz konstruiert und das Wirtszellspektrum dieses Virus untersucht. Die Reparatur des F11L-Gens ermöglichte MVA die Induktion von Zellbewegung nach Infektion, jedoch blieben seine unvollständige Morphogenese und eingeschränkte Vermehrungsfähigkeit in Säugetierzellen unbeeinflusst. F11L hat daher zumindest keine selbstständige Funktion als VACV host-range-Gen. Die Ergebnisse dieser Arbeit sind ein Beitrag zum besseren Verständnis der komplexen Virus-Wirts-Interaktionen des VACV sowie des eingeschränkten Wirtstropismus des Impfvirus MVA.
Untersuchung der Konformation und Dynamik von RNA mit Hilfe fluoreszierender Farbstoffmoleküle

Die vorliegende Arbeit beschäftigt sich mit der Untersuchung der konformationellen und elektronischen Eigenschaften sowie der Dynamik verschiedener RNA-Systeme. Zur Durchführung dieser Experimente wurde zusätzlich zu bereits vorhandenen statischen und zeitaufgelösten Absorptionsspektrometern im Rahmen dieser Arbeit eine Apparatur zur Messung von Fluoreszenzlebensdauern entwickelt, die durch die integrative Verwendung zweier verschiedener, etablierter Technologien (TCSPC und Aufkonvertierung) über einen weiten Zeitbereich von 9 Größenordnungen (100 fs - 0,1 ms) operiert. Mit diesem Aufbau konnten neben den RNA-Studien wichtige Beiträge zum Verständnis der Isomerisierung eines Retinalproteins, des Transportprozess des Membrantransportproteins TbSMR und der im Infraroten liegenden Fluoreszenz des Radikalkations von Astaxanthin gewonnen werden. Der Schwerpunkt der vorliegenden Arbeit liegt auf der Untersuchung verschiedener RNA-Systeme: So werden die optischen Eigenschaften einer 1-Ethinylpyren-modifzierten RNA-Adeninbase allein und in RNA-Strängen eingebunden untersucht. Statische Fluoreszenzmessungen zeigen einen ausgeprägten Ladungstransfercharakter des Chromophors und eine generell große Wechselwirkung zwischen Ethinylpyren und Adenin, die in einer substanziellen Änderung der optischen Eigenschaften des Pyrens resultiert. Die Untersuchung der schnellen Photodynamik von Pyrenadenin zeigt zudem eine Verringerung der Lebensdauer von Pyren um etwa 2 Größenordnungen. Pyrenadenin zeigt sowohl Fluoreszenz eines neutralen (100-200 ps), als auch eines energetisch tiefer liegenden Ladungstransferzustands (1-2 ns). Die Formationszeit des Ladungstransferzustandes fällt mit steigender Polarität des Lösemittels. Eingebunden in Modell-RNA-Stränge ist Fluoreszenzquantenausbeute des Chromophors ein deutlicher Indikator für seine Interkalation. Nur in der stabileren Umgebung von GC-Basenpaaren ist das Pyren in der Lage, sich dauerhaft innerhalb des Duplex aufzuhalten, während in einer flexibleren AU-Umgebung eine Position außerhalb des RNA-Duplex präferiert wird. Transiente Absorptionsmessungen zeigen, dass die Photophysik des in RNA eingebundenen Pyrenadenins nur kleine Variationen im Vergleich zur Photophysik des Labels allein aufweist. Die deutliche Abnahme der Quantenausbeute des interkalierten Chromophors geht hauptsächlich auf Kosten der langlebigeren Ladungstransferfluoreszenz, so dass interkaliertes Pyren insgesamt schneller in den Grundzustand zurückkehrt als nicht interkaliertes. Mit Hilfe eines doppelt modifizierten Duplex, bei dem sich jeweils ein Farbstoff an einem der beiden Stränge befindet, kann nachgewiesen werden, dass aufgrund von Exzimerwechselwirkungen eine Verschiebung des Fluoreszenzmaximums von 35 nm auftritt. Kurzzeitspektroskopische Messungen zeigen Signale, die als Superposition von Monomeren und Exzimeren interpretiert werden können, wobei die Lebensdauer des letzteren mit 18,5 ns die der Monomerkomponente um ein Vielfaches übertrifft. Ein weiterer Teil dieser Arbeit beschäftigt sich mit einer Studie zur Bindung des fluoreszenten Liganden Tetrazyklin an das Tetrazyklin bindende Aptamer. Hier wird auf Basis verschiedener Mutanten mit Hilfe des TCSPC eine Analyse der Stabilität der Bindetasche sowie mit der Stopped-Flow-Methode eine Beobachtung des Bindungsprozesses durchgeführt. Insgesamt folgt die Bindung des Tetrazyklins an das Aptamer einer zweistufigen Kinetik, deren zweiter Schritt irreversibel ist. Die Bindung läuft, verglichen mit anderen Aptameren, sehr schnell ab. Während die Mutationen von A13 und A50,die direkte Kontakte zum Substrat bilden, nur einen leichten Einfluss auf beide Bindungsschritte ausüben, führt eine Mutation der für die Präformation verantwortlichen Base A9 zu einer Verlangsamung des Bindungsprozesses um mehr als einen Faktor 20 durch eine immens gesteigerten Rückreaktionsrate des ersten Bindungsschritts. Hieraus lässt sich schließen, dass bei fehlender Präformation des Aptamers nur wenige Tetrazyklinmoleküle ein für vollständige Bindung geeignetes Aptamer vorfinden. Die Bindung an A13 und A50 geschieht bereits im ersten Schritt des Bindungsprozesses. Ferner konnte anhand von Lebensdauermessungen gezeigt werden, dass nach dem Wildtyp die Mutante A9G die stabilste Bindetasche aufwies. Das Fehlen eines direkten Kontaktes wirkt sich deutlich stärker aus. Insbesondere führt die Abwesenheit der Fixierung des Gegenions durch A50 zu der instabilsten Bindetasche. Wie in dieser Arbeit gezeigt wird, ist die zeitaufgelöste optische Spektroskopie insbesondere in Verbindung mit fluoreszierenden Molekülen ein ausgezeichnetes Mittel zur Beobachtung von Struktur und Dynamik von RNA. Die Empfindlichkeit von Fluoreszenz auf die Veränderung der Umgebung des Chromophors erlaubt es, Konformationsdynamik und elektronische Konfigurationen in Echtzeit zu beobachten.
Im Rahmen dieser Arbeit ist es gelungen, eine weltweit einmalige Messapparatur zu entwickeln, mit der Wasserstoffmolekülionen mittels kurzer Laserpulse ionisiert und die Reaktionsprodukte kinematisch vollständig vermessen werden können. Es wird dazu eine an die Coltrims-Technik angelehnte Detektionsmethode genutzt, bei der sowohl Protonen als auch Elektronen über den vollen Raumwinkel nachgewiesen werden können. Die H2+ -Ionen stammen aus einer Hochfrequenz-Ionenquelle und werden auf 400keV beschleunigt. Die Besetzungshäufigkeit der Vibrationsniveaus entspricht daher der Franck-Condon-Verteilung für den Übergang aus dem Grundzustand des neutralen Wasserstoffmoleküls in den elektronischen Grundzustand des Molekülions: H2 (xPg, ν = 0) → H2+ (1sσg, ν′) Dieser Ionenstrahl wird mit einem 780 nm Laserpuls der Pulslänge 40 fs überlappt. Nach der Reaktion fragmentiert das Molekülion entweder über den Dissoziationskanal H2+ + nhν ⇒ H + H+ oder über eine Ionisation gefolgt von einer Coulomb-Explosion: H2+ + nhν ⇒ H+ + H+ + e−. Die Projektile werden nach einer Driftstrecke von etwa 3 m auf einem Ionendetektor nachgewiesen. Für den Nachweis der Elektronen wurde ein spezielles Spektrometer konzipiert, das eine Unterdrückung ungewollter Elektronen erlaubt und so die Messung der Elektronen ermöglicht. Um Elektronen auszublenden, die vom Laser aus dem Restgas ionisiert werden, ist der Elektronendetektor in Flugrichtung der Ionen versetzt angebracht. Durch die unkonventionelle Ausrichtung des Lasers in einem Winkel von 20◦ relativ zur Flugrichtung der Ionen können vom Laser erzeugte Elektronen nur dann den Elektronendetektor erreichen, wenn sie aus dem bewegten Bezugsystem der Projektile stammen. Diese Unterdrückung macht die Messung der Elektronen erst möglich, hat aber auch eine nachteilige Geometrie der Verteilungen gegenüber den Detektorebenen zur Folge. Durch die Ausnutzung der Projektilgeschwindigkeit ist überdies die Benutzung eines B-Feldes zur Verbesserung der Flugzeitauflösung der Elektronen nicht möglich. Um eine Überlappung des Ionenstrahls mit dem Laserfokus zu erreichen, wurde im Bereich der Reaktionszone ein System zur Visualisierung der Strahlpositionen integriert. Dieses kann überdies für eine Intensitätseichung bei linear polarisiertem Licht verwendet werden. Bei der Reaktion kommt es durch die vergleichsweise lange Pulsdauer schon bei relativ niedrigen Intensitäten zu Dissoziationsprozessen. Das dissoziierende Molekül erreicht noch während der ansteigenden Flanke des Laserpulses auf diese Weise Abstände, bei denen der Prozess der Charge-Resonance-Enhanced-Ionization (CREI) stattfinden kann. Auch die in einem sehr engen Winkelbereich um die Polarisationsrichtung des Lasers liegende Winkelverteilung der gemessenen Protonen deutet darauf hin, dass CREI der dominante Ionisationsprozess ist. Durch die vorausgehende Dissoziation nimmt das Molekül schon vor der Ionisation eine kinetische Energie auf, so dass die gemessene KER-Verteilung einer Summe aus KERDissoziation und KERIonisation darstellt. Ein Vergleich mit den KER-Spektren des Dissoziationsprozesses zeigt, dass die aufgenommene Energie durch Dissoziation zu einem überwiegenden Anteil in einem Bereich von 0, 6 ± 0, 35 eV besitzt, während die Gesamt-KER-Verteilung deutlich höhere Werte bis zu 6 eV aufweist. Dies ermöglicht, aus der gemessenen KER-Verteilung den internuklearen Abstand zum Ionisationszeitpunkt näherungsweise zu bestimmen. Die gemessenen Elektronen weisen, ebenso wie die Protonen, eine scharfe Ausrichtung entlang der Laserpolarisation auf, was durch den Einfluss des Lasers auf dieser Achse nicht verwunderlich ist. Bei zirkularer Polarisation dagegen findet eine Netto-Beschleunigung der Elektronen senkrecht zur Richtung des elektrischen Feldes zum Ionisationszeitpunkt statt, sodass die Messung der Elektronenimpulse eine geeignete Messgröße zur Untersuchung des Ionisationsprozesses darstellt. Auf diese Art konnten Winkelverteilungen der Elektronen bezüglich der internuklearen Achse innerhalb der Polarisationsebene gemessen werden. Abhängig von KER und Elektronenergie konnte dabei eine Verdrehung der Verteilung gegenüber den klassisch erwarteten 90◦ relativ zur internuklearen Achse festgestellt werden. Die Winkelverteilung rotiert dabei mit steigendem KER entgegen des Drehsinns. Dies widerspricht der gängigen Vorstellung einer Tunnelionisation, bei der nur die Beschleunigung des Elektrons im Laserfeld eine Rolle spielt und der Einfluss des Coulomb-Potentials vernachlässigt wird. Für höhere Elektronenergien zeigt sich eine zweite konkurrierende Struktur, die für die höchsten Energien die sonst vorherrschende erste Struktur sogar dominiert. Da sich in den Protonenspektren für linear polarisiertes Licht kein Einfluss einer Ionisationsenkrecht zur Polarisationsrichtung findet, erscheint dies als Grund für die zweite Struktur in den Elektron-Winkelverteilungen als unwahrscheinlich. Eine stichhaltige und gestützte Erklärung gibt es bisher weder für die Rotation der ersten Struktur noch für die Herkunft der zweiten. Dies zeigt deutlich, dass es auch für dieses einfachste Molekülsystem noch einen erheblich Handlungsbedarf sowohl auf theoretischer als auch von experimenteller Seite gibt. Da dieses Experiment den ersten experimentellen Zugang für die direkte Untersuchung der Elektronimpulse bei der Ionisation von H2+ -Ionen in kurzen Laserpulsen darstellt, bietet sich hier die bisher einzige Möglichkeit, dieses Verhalten experimentell zu untersuchen.
Die Wirkungen von Acrylamid beschäftigen seit Jahrzehnten Mediziner, Toxikologen, Biologen und Analytiker. Zwar konnte eine kanzerogene Wirkung beim Tier eindeutig nachgewiesen werden, der Nachweis dieser Wirkung beim Menschen ist aber bis heute nicht erbracht. Bis 2000 wurde angenommen, dass Acrylamid ausschließlich technisch erzeugt vorkommt und der Mensch Acrylamid nur aus dieser Quelle aufnimmt. Hierauf beruhen Grenz- und Richtwerte auf nationaler und internationaler Ebene, sowohl am Arbeitsplatz als auch für Trinkwasser und Verpackungsmaterialien. Neben dem Acrylamidgehalt in technischen Produkten, Wasser und Luft gelang der Nachweis der aufgenommenen Acrylamidmenge über Acrylamid-Hämoglobin-Addukte im Blut erst 1995, nachdem der Acrylamid-Metabolismus aufgeklärt worden war und die analytischen Methoden verfeinert waren. Die Annahme, dass Acrylamid ausschließlich in technischen Prozessen gebildet wird, war jedoch nicht vereinbar mit Erkenntnissen, dass auch Probanden einen erhöhten Acrylamidspiegel im Blut aufzeigten, die nicht mit Acrylamid am Arbeitsplatz in Kontakt gekommen waren. Im Jahr 2002 gelang es eindeutig nachzuweisen, dass Acrylamid in vielen, meist alltäglich verzehrten Lebensmitteln abhängig von der Zubereitungsart zu finden ist. Mit Hilfe einer verbesserten Analytik konnten schließlich hohe Acrylamidmengen in zahlreichen, vor allem frittierten Lebensmitteln, belegt werden, die die Belastung aus technischen Quellen oft weit überschritten. Die Tatsache, dass die Nahrung der allgemeinen Bevölkerung deutlich Acrylamid-belastet ist, hat die Bewertung der Acrylamid-Toxizität und die Maßnahmen zum Gesundheitsschutz nachhaltig verändert. Durch den späten Nachweis der Acrylamid-Aufnahme mit Lebensmitteln sind viele frühere Annahmen für die Gesundheitsbewertung irrig, die hieraus gezogenen Schlußfolgerungen nicht haltbar. Seit 2002 ist die Reduktion der Lebensmittelbelastung in den Mittelpunkt gerückt, die Arbeitsplatzbelastung hat - mit Ausnahme der Schleimhautreizung - keine weitere Berücksichtigung erhalten. Die hohe Belastung von Lebensmitteln stellt bisherige Richtlinien und Expositionsauflagen in Frage, der fehlende bisherige epidemiologische Nachweis, der aus Tierversuchen eindeutig nachweisbaren Karzinogenität stellt für Acrylamid auch die bisherigen Modell-Annahmen der Krebsentstehung durch Umweltschadstoffe in der „one hit-Hypothese“ in Frage.
Die chronische Hepatitis C Virus-Infektion ist eine der häufigsten Ursachen für Leberzirrhose und das hepatozelluläre Karzinom. CD81 ist ein Oberflächenprotein aus der Familie der Tetraspanine, welches auf Hepatozyten und Lymphozyten exprimiert ist. CD81 interagiert mit dem Hepatitis C Virus-Hüllprotein E2. Die Interaktion ist essentiell für die Infektion von Hepatozyten durch das Hepatitis C Virus und beteiligt an der Hepatitis C Virus assoziierten Immunmodulation, welche in Zusammenhang mit der Chronifizierung und dem Auftreten von autoimmunologischen Phänomenen steht. Verschiedene Untersuchungen haben gezeigt, dass CD81 von Lymphozyten sezerniert werden kann. Die Bedeutung der CD81-Serumkonzentration wurde bislang nicht untersucht. Um neue Erkenntnisse über die Rolle von CD81 innerhalb der Pathogenese der chronischen Hepatitis C zu erlangen, wurde in der vorliegenden Arbeit die CD81-Serumkonzentration bei Patienten mit chronischer Hepatitis C untersucht. Nach Anreicherung von CD81 mittels differentieller Zentrifugation konnte durch Anwendung des Western Blot-Verfahrens gezeigt werden, dass CD81 im Serum bei Patienten mit chronischer Hepatitis C im Vergleich zu gesunden Probanden signifikant erhöht ist (p = 0,001). Im Hinblick auf die CD81-vermittelte Immunmodulation wurde anschließend die CD81-Serumkonzentration mit der Höhe der Lebertransaminase GPT als Surrogatmarker für die entzündliche Aktivität der Leber verglichen. Die Untersuchungen ergaben eine signifikante Korrelation zwischen der CD81- Serumkonzentration und der Höhe der GPT (Korrelationskoeffizient r = 0,334, p = 0,016). Im nächsten Schritt wurde der Einfluss der antiviralen Therapie auf die CD81- Serumkonzentration untersucht. Bei Patienten mit chronischer Hepatitis C und erhöhten Transaminasen, die nach Therapie ein dauerhaftes Ansprechen entwickelten, zeigte sich unter Therapie ein signifikanter Abfall der CD81-Serumkonzentration im Vergleich zu der vor Therapie (p = 0,045). Zusammenfassend zeigen diese Ergebnisse der Promotionsarbeit, dass CD81 im Serum nachweisbar ist, dass die CD81-Serumkonzentration bei Patienten mit chronischer Hepatitis C erhöht ist und dass es einen Zusammenhang zwischen Leberentzündung und Therapieansprechen gibt. Diese Ergebnisse deuten daraufhin, dass CD81 für die Pathogenese der Hepatitis C Virus-Infektion möglicherweise von Bedeutung ist.
Zielstellung: Die Untersuchung des klinischen Effekts der topischen subgingivalen Applikation eines 14%igen Doxycyclin-Gels zusätzlich zu mechanischem Debridement (DEB) während der unterstützenden Parodontitistherapie (UPT) auf die Furkationsbeteiligung mehrwurzeliger Zähne. Material und Methoden: Bei 39 UPT-Patienten mit zumindest vier Zähne mit Sondierungstiefen (ST) ≥ 5 mm und Bluten nach Sondieren (BOP) (davon mindestens eine Furkationsstelle) erfolgte ein DEB aller Stellen mit ST ≥ 4mm. Nach Randomisierung wurde bei 19 Patienten zusätzlich ein 14%iges Doxycyclin-Gel subgingival instilliert (DEB & DOXY). Die klinischen Parameter wurden vor der Therapie (Baseline) sowie 3, 6 und 12 Monate danach erhoben. Der zusätzliche Nutzen des Doxycyclin-Gels wurde bestimmt als 1) Verbesserung der Furkationsbeteiligung und 2) Einfluss auf die Häufigkeit der Reinstrumentierung während der 12 Monate. Ergebnisse: Insgesamt wurden 323 Furkationsstellen (Grad 0: 160; Grad I: 101; Grad II: 18; Grad III: 44) behandelt (DEB: 165; DEB & DOXY: 158). DEB & DOXY resultierte nach 3 Monaten in einer stärkeren Reduktion der Furkationsbeteiligung als alleiniges DEB (p = 0,041). Allerdings führte DEB & DOXY zu keiner Reduktion der Häufigkeit der Reinstrumentierung während der 12 Monate. Schlussfolgerung: Die topische subgingivale Doxycyclingabe zusätzlich zu DEB hat einen kurzzeitigen Effekt auf die Furkationsbeteiligung. Aber es reduziert nicht die Häufigkeit der Reinstrumentierung über einen Zeitraum von 12 Monaten.
Der Soziologe Johannes TWARDELLA analysiert in seinem Buch "Pädagogischer Pessimismus" den vollständigen Verlauf einer Unterrichtsstunde im Fach Deutsch in der 10. Jahrgangsstufe einer Hauptschule. Aus einem "wunderschönen guten Morgen" – so beginnt das Transkript – wird eine kleine Katastrophe. Wie konnte das passieren? Die detaillierte Analyse TWARDELLAs zeigt eindrücklich, dass das Verhältnis der Lehrkraft zu den Schülerinnen und Schülern sowie zu ihrer Profession gestört und widersprüchlich ist. Auf der einen Seite ist der Unterricht geprägt von einer negativen Anthropologie des Schülers bzw. der Schülerin, dem pädagogischen Pessimismus. Auf der anderen Seite besteht aus Sicht der Lehrkraft der optimistische Glaube an die didaktische Lösung des handlungsorientierten Unterrichts. Letztlich wird erkennbar, dass sich durch eine Self-Fulfilling Prophecy diese abgründige Kombination zu einer veränderungsresistenten Ideologie verhärtet und am Ende nur noch die Aufrechterhaltung des Betriebs steht – so sinnfrei er auch geworden sein mag. Das vorliegende Buch wird in den Kontext der derzeitigen bildungspolitischen und bildungswissenschaftlichen Diskussion gesetzt.
Unternehmen und Ethik

Die noch nicht völlig überwundene Finanzmarktkrise hat nicht nur den Gesetzgeber auf den Plan gerufen. Auch die Frage nach der Ethik der Akteure wird vielfach erörtert. Haben von Gier getriebene Finanzmarktakteure nicht nur Rechtsregeln, sondern auch ethische Normen, die Grundsätze des Wirtschaftens ehrbarer Kaufleute, gewissenlos beiseite geschoben, um sich zu bereichern? Wie läßt sich die Beachtung dieser Normen künftig sichern? Diese aktuelle Debatte soll Anlaß zu einigen allgemeineren Betrachtungen zum Thema „Unternehmen und Ethik“ sein.
In dieser Arbeit geht es um eine theaterwissenschaftliche Analyse ausgewählter Stücke des Theaterregisseurs und Autors René Pollesch. Bei den Stücken handelt es sich um die Heidi Hoh-Trilogie (1999-2001), Insourcing des Zuhause (2001), Der Leopard von Singapur (2003) und Cappuccetto Rosso (2005). Die Sujets und Stilmerkmale des Theaters von Pollesch werden dabei mittels der Theorien des Philosophen Jacques Rancière als Elemente eines politischen Theaters interpretiert, dem es wesentlich darum geht, alle Formen der Regelhaftigkeit, der symbolischen Ordnung oder der etablierten Sichtbarkeit durch Unterbrechung der Regel, Aufhebung der Ordnung, Sichtbarmachung des Unsichtbaren oder Verstimmlichen des Stimmlosen, aber auch durch Formen einer subversiven Wiederholung und Aneignung kritisierter Sicht- und Sprechweisen zu provozieren. Um das Politische bei Pollesch zu erkunden, werden Rancières Theorietexte „angewandt", indem sie während der analytischen Betrachtung der Pollesch-Abende in ästhetische Strategien umgewandelt werden. Dies geschieht auf drei Ebenen: 1: Darstellungsform, 2: Arbeitsweise, 3: Die auf der Bühne sichtbar agierenden Subjekte. Rancière erscheint mit seinem Konzept einer gegebenen gesellschaftlichen Ordnung als „Aufteilung des Sinnlichen" ideal geeignet, um das Politische auf eben diese drei genannten Ebenen aufzuspüren. Sein Modell von Politik, so zeigt diese Arbeit, erlaubt damit die ästhetische Praxis René Polleschs auf verschiedenen Ebenen als politisch zu kennzeichnen, aber es führt auch auf einige Aporien, denen diese Praxis gerade in ihrem politischen Anspruch ausgeliefert ist.
Die Augsburger historische Dissertation von Andreas Toppe ist aus dem Projekt "Wehrmacht in der nationalsozialistischen Diktatur" des Münchner Instituts für Zeitgeschichte hervorgegangen. In der Einleitung legitimiert sich der Verfasser durch die gesellschaftlichen Diskussionen, welche im Zusammenhang mit der sogenannten Wehrmachtsausstellung Mitte der 1990er Jahre entstanden. Sie hätten gezeigt, so Toppe, dass "die deutschen Kriegsverbrechen hauptsächlich nach den moralischen Kategorien der'Nachgeborenen' bewertet werden, nicht aber nach den damals herrschenden Maßstäben des international gültigen Kriegs- und Völkerrechts" (9). Das Buch will demnach zu einer Verrechtlichung des Werturteils beitragen, diese Verrechtlichung wiederum soll dann eine "sachlich korrekte Verortung der Wehrmacht in der Geschichte" ermöglichen, an der es aus Sicht des Verfassers wohl fehlt; klar gesagt wird dies jedoch nicht. ...
Vibronic (vibrational-electronic) transition is one of the fundamental processes in molecular physics. Indeed, vibronic transition is essential both in radiative and nonradiative photophysical or photochemical properties of molecules such as absorption, emission, Raman scattering, circular dichroism, electron transfer, internal conversion, etc. A detailed understanding of these transitions in varying systems, especially for (large) biomolecules, is thus of particular interest. Describing vibronic transitions in polyatomic systems with hundreds of atoms is, however, a difficult task due to the large number of coupled degrees of freedom. Even within the relatively crude harmonic approximation, such as for Born-Oppenheimer harmonic potential energy surfaces, the brute-force evaluation of Franck-Condon intensity profiles in a time-independent sum-over-states approach is prohibitive for complex systems owing to the vast number of multi-dimensional Franck-Condon integrals. The main goal of this thesis is to describe a variety of molecular vibronic transitions, with special focus on the development of approaches that are applicable to extended molecular systems. We use various representations of Fermi’s golden rule in frequency, time and phase spaces via coherent states to reduce the computational complexity. Although each representation has benefits and shortcomings in its evaluation, they complement each other. Peak assignment of a spectrum can be made directly after calculation in the frequency domain but this sum-over-states route is usually slow. In contrast, computation is considerably faster in the time domain with Fourier transformation but the peak assignment is not directly available. The representation in phase space does not immediately provide physically-meaningful quantities but it can link frequency and time domains. This has been applied to, herein, for example (non-Condon) absorption spectra of benzene and electron transfer of bacteriochlorophyll in the photosynthetic reaction center at finite temperature. This work is a significant step in the treatment of vibronic structure, allowing for the accurate and efficient treatment of complex systems, and provides a new analysis tool for molecular science.
In this Letter, we propose a new scenario emerging from the conjectured presence of a minimal length ℓ in the spacetime fabric, on the one side, and the existence of a new scale invariant, continuous mass spectrum, of un-particles on the other side. We introduce the concept of un-spectral dimension DU of a d-dimensional, euclidean (quantum) spacetime, as the spectral dimension measured by an “un-particle” probe. We find a general expression for the un-spectral dimension DU labelling different spacetime phases: a semi-classical phase, where ordinary spectral dimension gets contribution from the scaling dimension dU of the un-particle probe; a critical “Planckian phase”, where four-dimensional spacetime can be effectively considered two-dimensional when dU=1; a “Trans-Planckian phase”, which is accessible to un-particle probes only, where spacetime as we currently understand it looses its physical meaning.
Die akustische Mikroskopie stellt ein neuartiges bildgebendes Verfahren zur zerstörungsfreien Werkstoffprüfung dar, das erst seit kurzem in der Medizin eingesetzt wird und bei dem reflektierte Ultraschallwellen zur Bilderzeugung verwendet werden. Auf diese Weise können neue Informationen bezüglich der Materialparameter des untersuchten Gewebes erlangt werden. Neben einer enorm großen Bandbreite an möglichen Auflösungen bietet diese Technik die Möglichkeit, opake Objekte zu durchdringen und ihr Inneres abzubilden. Durch seine nicht-destruktive Art und die Notwendigkeit einer Kopplungsflüssigkeit bietet diese mikroskopische Methode zudem die Option lebende Zellen und Gewebe zu untersuchen. Das Ziel der durchgeführten Arbeit war es, die Interpretation von akustischen Bildern des menschlichen Gewebes zu erleichtern. Zu diesem Zweck analysierten wir SAM-Abbildungen von Knochengewebe im Vergleich zu anderen Untersuchungsverfahren. Neben einer qualitativen Bewertung der Morphologie gleichartiger Strukturen wurden ausgedehnte morphometrische Messungen und energiedispersive Röntgenanalysen durchgeführt. Als Methoden für den qualitativen Vergleich wurden konventionelle mikroskopische Techniken wie Polarisations-, Fluoreszenz-, Durchlicht-, Auflicht- und Rasterelektronenmikroskopie sowie Mikroradiographie vergleichend verwendet. Der morphometrische Vergleich erfolgte zwischen ultraschall- und auflichtmikroskopisch erzeugten Abbildungen. Weiterhin wurde die Beziehung zwischen akustischen Grauwertdarstelllungen und energiedispersiven Röntgenanalysen bewertet. Als Untersuchungsmaterial dienten Mandibulaknochen von sieben Beagle-Hunden. Die Mandibula eines jeden Hundes wurde beidseits mittels Osteoskalpell nach Sachse (Bien Air, CH-Biel) und CO2-Laser (entwickelt im Center of Advanced European Studies and Research, D-Bonn) am Margo ventralis im unbezahnten Kieferabschnitt osteotomiert. Nach einer dreiwöchigen Heilungsphase wurden die Knochen entnommen und vor der Schnittfertigung in Polymethylmethacrylat eingebettet. Beim qualitativen Vergleich der Morphologie gleichartiger Strukturen fanden sich bei SAM-Abbildungen und bei Aufnahmen, die mit den anderen mikroskopischen Techniken erzeugt wurden, weitgehende Übereinstimmungen hinsichtlich gröberer morphologischer Merkmale wie Umriss, flächiger Ausdehnung und Lage von Perforationen. Diese Gemeinsamkeiten konnten sowohl an Kallusgewebe als auch an osteonalem Knochen beobachtet werden. Unterschiede fanden sich hinsichtlich feinerer morphologischer Merkmale wie der geometrischen Übereinstimmung im Detail und der flächigen Ausdehnung bei höherer Vergrößerung. Zudem stellte sich die Binnenstruktur von Kallus und osteonalem Knochengewebe differenzierter dar als mit Hilfe der anderen mikroskopischen Techniken. Morphometrische Messungen zeigten, dass Strukturen, die im Bereich des Osteotomiespalts gemessen worden waren, im Ultraschallbild tendenziell größer abgebildet werden als in Auflichtaufnahmen. Im Gegensatz dazu wurden Strukturen im Bereich des kortikalen Knochens im ultraschallmikroskopischen Bild eher unterschätzt. Diese Ungenauigkeit bei der Abbildung von Strukturen im Ultraschallbild lässt sich am ehesten dadurch erklären, dass durch unterschiedliche Eindringtiefen des Ultraschalls auch unterschiedlich dicke Ebenen der Probe abgebildet werden. Wird eine dickere Schicht der Probe durch das akustische Mikroskop dargestellt als durch das Auflichtmikroskop, so kann es zu einer Überschätzung der Strukturen kommen und umgekehrt. Anhand energiedispersiver Röntgenanalysen konnte ebenfalls gezeigt werden, dass die Zunahme des Calcium- bzw. des Phosphorgehalts nicht alleine eine Aufhellung der Grautöne im Ultraschallbild verursachen kann. Auch der Vergleich mit Abbildungen der Elektronenmikroskopie ließ Unterschiede der Grauwerte erkennen. Demzufolge wird das akustische Bild nicht nur durch den Mineralgehalt, die Dichte und die atomare Zusammensetzung des Knochengewebes beeinflusst. Damit konnte mit der vorliegenden Untersuchung nachgewiesen werden, dass die Ultraschallmikroskopie ein neuartiges Verfahren darstellt, mit dem sich Knochen nicht nur in seiner Mikrostruktur morphologisch darstellen lässt, sondern mit der sich auch neuartige Informationen über funktionelle Parameter auf Nanoebene gewinnen lassen. Die Bedeutung dieser enormen Möglichkeiten sind im Moment noch nicht abschätzbar, weitere Untersuchungen müssen folgen.
The first measurement of two-pion Bose–Einstein correlations in central Pb–Pb collisions at √sNN=2.76 TeV at the Large Hadron Collider is presented. We observe a growing trend with energy now not only for the longitudinal and the outward but also for the sideward pion source radius. The pion homogeneity volume and the decoupling time are significantly larger than those measured at RHIC.
The mass-dependent structure of the composite nucleus is shown based on three-dimensional timedependent Hartree-Fock calculations with Skyrme interactions (SLy4d and SkM*). One remarkable result is that the isovector monopole excitation dominantly appears for collisions of heavy nuclei, and the isovector dipole excitation for those of light ones. Such a difference found in the dynamical structure of composite nucleus plays a role in the equilibration of charge.
Methodik und Material: Injektion von Kolonkarzinomzellen der Zellreihe CC531 subkapsulär in den linken Leberlappen bei 36 männlichen WAG-Ratten (Gewicht 180-220g). Induziert wurden zwei unterschiedlich starke Tumorkonzentrationen: Gruppe A (n = 18, Injektion von 1 Mio. Tumorzellen) und Gruppe B (n = 18, Injektion von 4 Mio. Tumorzellen). Messung der Tiere nach 14 Tagen am CT Siemens Somatom Sensation 64 und Analyse des Tumorvolumens mit dem Volume Viewer Plus von Centricity AW Suite. Unterschiedlich starkes Wachstum der Lebertumore führte zu einer Einteilung der Ratten in Gruppe A(1,2) (n = 12): Tumorvolumen <3,5cm3 und Gruppe B(1,2) (n = 12): Tumorvolumen ≥3,5cm3. Die volumenbezogene Therapie erfolgte an Tag 15 wie folgt: Bei jeweils 6 Tieren der Gruppen A1 und 6 Tieren der Gruppe B1 wurde eine systemische Chemotherapie mit 0,1mg/ml Mitomycin C (Standardtherapie) durchgeführt. Die andere Hälfte der Tiere (6 Tiere der Gruppe A2 und 6 Tiere der Gruppe B2) erhielten die doppelte Konzentration von 0,2mg/ml (< 10% der letalen Dosis). 6 Tiere dienten der pathologischen Auswertung, weitere 6 Tiere dienten als Kontrollgruppe. An Tag 25 erfolgte eine erneute Bestimmung des Tumorvolumens. Ergebnisse: Die Therapie wurde von allen Ratten gut toleriert. Das durchschnittliche Tumorvolumen betrug in A (1,2) 2,18cm3 +/- 0,82 (range: 0,55 – 3,42) und in Gruppe B (1,2) 4,52cm3 +/- 0,82 (range: 3,72 – 5,86). Das CT-follow up an Tag 25 ergab, dass eine erhöhte Mitomycinkonzentration zu einer Tumorregression von 20,55 % in der Gruppe A2, 21,03 % in B1 und 46,61% in B2 führte. In der Gruppe A1 zeigte sich ein Progress des Tumorvolumens von 51,38%. Schlussfolgerung: Chemotherapeutika werden in Abhängigkeit von Körpergewicht, Körpergröße und Körperoberfläche verabreicht. Einen neuartigen Ansatz stellt die tumorvolumenadaptierte Chemotherapie (TVACT) mit Mitomycin C dar, da große Tumorvolumen eine bessere Ansprechbarkeit auf Mitomycin C zeigen als kleinere. Die TVACT ist somit eine neue relevante Behandlungsoption bei der Behandlung von malignen Tumoren im Tiermodell. Hinsichtlich der wachsenden Bedeutung neuer Staging Systeme im avanzierten kolorektalen Karzinom sollte das Tumorvolumen auch als prognostischer Faktor berücksichtigt werden.
The inclusive charged particle transverse momentum distribution is measured in proton–proton collisions at s=900 GeV at the LHC using the ALICE detector. The measurement is performed in the central pseudorapidity region (|η|<0.8) over the transverse momentum range 0.15<pT<10 GeV/c. The correlation between transverse momentum and particle multiplicity is also studied. Results are presented for inelastic (INEL) and non-single-diffractive (NSD) events. The average transverse momentum for |η|<0.8 is 〈pT〉INEL=0.483±0.001 (stat.)±0.007 (syst.) GeV/c and 〈pT〉NSD=0.489±0.001 (stat.)±0.007 (syst.) GeV/c, respectively. The data exhibit a slightly larger 〈pT〉 than measurements in wider pseudorapidity intervals. The results are compared to simulations with the Monte Carlo event generators PYTHIA and PHOJET.
In dieser Studie konnte deutlich gezeigt werden, dass TS eine größere psychische Belastung vorweisen, als die Norm. Sie neigen stärker zur Somatisierung und sind insgesamt zwanghafter, ängstlicher,aggressiver und depressiver. Dies spricht für eine große psychische Belastung, die am ehesten von den schwierigen Begleitumständen und der enormen Bandbreite einer Geschlechtsidentitätsstörung herkommen. Durch die Reaktionen im privaten und beruflichen Umfeld verändern sich bodenständige Strukturen, wie Verhältnis zu den Eltern, Partnerschaftsverhältnisse und Freundschaften. Ein vermindertes Selbstwertgefühl entsteht konsekutiv aus der sozialen Ausgrenzung und dem langen Verlauf zwischen Diagnosestellung und Therapie mit einem Endergebnis. Das verminderte Selbstwertgefühl spielt eine große Rolle im Hinblick auf die Therapie. Denn 50 % der TM und 100 % der TF korrelieren die Geschlechtsidentität mit dem Äußeren und streben deshalb als Therapieziel eine Geschlechtsangleichung an. Dass in diesen Fällen das Selbstwertgefühl durch eine operative Geschlechtsangleichung gesteigert und deutlich verbessert werden kann konnte jedoch in dieser Studie nicht bewiesen werden. Ob das nun mit oft unzureichenden Operationsergebnissen zusammenhängt bleibt zunächst offen. Green und Blanchard 2000; van Kesteren, Gooren und Megens 1996 belegten bereits, dass nicht alle TS eine Geschlechtsangleichung anstreben. Somit sind diese Ergebnisse relativ konform mit den Ergebnissen dieser Studie. Eine weitere Rolle spielt das elterliche Erziehungs- und Beziehungsverhalten. Die meisten TS haben sowohl zum Vater als auch zur Mutter konflikthafte Beziehungen. Wobei in dieser Studie vor allem imponierte, dass TF ein konflikthafteres Verhältnis vor allem zur Mutter haben und weniger emotionale Wärme erfahren haben. Ob die Geschlechtsidentitätsstörung ursächlich für die konflikthaften Elternbeziehungen ist oder die konflikthafte Elternbeziehung erst durch ein Outcome der TS entstanden ist konnte nicht hinreichend geklärt werden, wäre aber für die Frage der elterlichen Erziehung als Ursache der TS sehr interessant. Des Weiteren konnte diese Studie zeigen, dass es zwischen TF und TM nur einen signifikanten Unterschied gibt und zwar, dass TF weniger emotionale Wärme durch die Mutter erfahren haben. Ansonsten unterscheiden sich TF von TM nicht wesentlich in punkto psychische Belastung, Selbstwertgefühl und Einfluss der elterlichen Beziehung auf die GIS. Weiterhin konnte in dieser Studie belegt werden, dass sich bereits operierte TS von noch nicht operierten TS nicht unterscheiden. Auch hier zeigt sich kein Unterschied im Hinblick auf die psychische Belastung und das Selbstwertgefühl bei bereits noch nicht operierten TS im Vergleich zu bereits operierten. Das Grundleiden bleibt das Gleiche, offen bleibt zu diskutieren, ob ein unzureichendes Operationsergebnis daran Schuld trägt oder die Tatsache, dass viele TS sich nicht nur zu EINEM Geschlecht zugehörig fühlen. Diese Tatsache konnte in dieser Arbeit eruiert werden und sollte weiterhin ein Diskussionspunkt hinsichtlich voreiliger geschlechtsangleichender Therapiemaßnahmen sein. Es gelang bedauerlicherweise nicht signifikante Korrelate/Risikofaktoren in direktem Bezug auf TS zu ermitteln. Die ermittelten signifikanten Korrelate sind nicht spezifisch für das Kollektiv der TS. Somit sind mangelndes Selbstwertgefühl und Attraktivitätsempfinden, sowie zunehmende Unsicherheit und Besorgnis auch begleitende Faktoren in anderen Patientenkollektiven (Tumorpatienten nach OP und Chemo, Brandopfer etc.). Für viele TS wäre es erstrebenswert nicht als „anders“ oder „krank“ gesehen zu werden, um ein höheres Ansehen in unserer Gesellschaft zu gewinnen. Jedoch gäbe es gar keine Unterschiede zu TS, dann würde vielen TS die Behandlungsgrundlage, nämlich eine Geschlechtsangleichung, genommen werden. Wenn auch nicht alle TS an einer Geschlechtsangleichung interessiert sind, gibt es wiederum TS, die auf die Diagnose Transsexualität angewiesen sind. Es gibt keine Diagnose ohne Krankheit und umgekehrt. Standardisierte Persönlichkeitstests sind nicht auf den individuellen Einzelfall zugeschnitten, sondern erfassen allgemeine, generalisierte Parameter. Jeder Fall von Transsexualität ist ein individueller Einzelfall. Ergebnisse eines Standardtests werden sich somit nicht pauschalisieren lassen. Die in o.g. Veröffentlichungen gemachten Angaben zu Transsexuellen sind durch das Sammeln vieler Einzelfälle in einem Zeitraum von mehreren Jahren entstanden (Balint 1960 und Becker 2003). Auf die Schnelle lässt sich da kein Psychopathologisches Profil erstellen. Viele Patienten befinden sich jahrelang vor und nach Geschlechtsangleichenden Maßnahmen in psychologischer Betreuung. Die psychologische Betreuung ist und bleibt somit das non plus ultra im Hinblick auf Diagnostik, Mit-Therapie und eigentlich fast lebenslange Begleitung. A comprehensive evaluation of seasonal backward trajectories initialized in the Northern Hemisphere lowermost stratosphere (LMS) has been performed to investigate the origin of air parcels and the main mechanisms determining characteristic structures in H2O and CO within the LMS. In particular we explain the fundamental role of the transit time since last tropopause crossing (tTST) for the chemical structure of the LMS as well as the feature of the extra-tropical tropopause transition layer (ExTL) as identified from CO profiles. The distribution of H2O in the background LMS above Θ=320 K and 340 K in northern winter and summer, respectively, is found to be governed mainly by the saturation mixing ratio, which in turn is determined by the Lagrangian Cold Point (LCP) encountered by each trajectory. Most of the backward trajectories from this region in the LMS experienced their LCP in the tropics and sub-tropics. The transit time since crossing the tropopause from the troposphere to the stratosphere (tTST) is independent of the H2O value of the air parcel. TST often occurs 20 days after trajectories have encountered their LCP. CO, on the other hand, depends strongly on tTST due to its finite lifetime. The ExTL as identified from CO measurements is then explained as a layer of air just above the tropopause, which on average encountered TST fairly recently.
A comprehensive evaluation of seasonal backward trajectories initialized in the northern hemisphere lowermost stratosphere (LMS) has been performed to investigate the factors that determine the temporal and spatial structure of troposphere-to-stratosphere-transport (TST) and it’s impact on the LMS. In particular we explain the fundamental role of the transit time since last TST (tTST) for the chemical composition of the LMS. According to our results the structure of the LMS can be characterized by a layer with tTST<40 days forming a narrow band around the local tropopause. This layer extends about 30K above the local dynamical tropopause, corresponding to the extratropical tropopause transition layer (ExTL) as identified by CO. The LMS beyond this layer shows a relatively well defined separation as marked by an aprupt transition to longer tTST indicating less frequent mixing and a smaller fraction of tropospheric air. Thus the LMS constitutes a region of two well defined regimes of tropospheric influence. These can be characterized mainly by different transport times from the troposphere and different fractions of tropospheric air. Carbon monoxide (CO) mirrors this structure of tTST due to it’s finite lifetime on the order of three months. Water vapour isopleths, on the other hand, do not uniquely indicate TST and are independent of tTST, but are determined by the Lagrangian Cold Point (LCP) of air parcels. Most of the backward trajectories from the LMS experienced their LCP in the tropics and sub-tropics, and TST often occurs 20 days after trajectories have encountered their LCP. Therefore, ExTL properties deduced from CO and H2O provide totally different informations on transport and particular TST for the LMS.
The TTL is the transition layer between the tropical troposphere and stratosphere, and is the main region where tropospheric air enters the stratosphere. In this thesis different transport processes are studied by using in situ measurements of tracers. Long-lived tracers were measured with the High Altitude Gas Analyzer (HAGAR) on board the M55 Geophysica aircraft. The instrument was developed by the University of Frankfurt and measures the long-lived tracers CO2, N2O, CFC-12, CFC-11, H-1211, SF6, CH4 and H2 with two gas chromatographic channels and a CO2 sensor (LICOR). The measurements are supported by CO and O3 measurements of other instruments. Two campaigns were conducted to obtain measurements in the TTL: SCOUT-O3 (November/December 2005 in Darwin, Australia) and AMMA-SCOUT-O3 (August 2006 in Ouagadougou, Burkina Faso). After a general introduction of the thesis in chapters one and two, the third chapter describes the findings during this last campaign. Five local flights are analyzed to study the different transport processes that occur in the tropical tropopause layer above West-Africa: deep convection up to the level of main convective outflow, vertical mixing after overshooting of air in deep convection, horizontal inmixing from the extratropical lower stratosphere, and horizontal transport across the subtropical barrier. Main findings are that the TTL over West-Africa is mostly influenced by remote convection. The subtropical barrier is not a strong barrier but more a region of transition between the extratropical and the tropical stratosphere. Chapter 4 presents the results obtained during the SCOUT-O3 campaign. From the eight local flights the last four flights (051129, 051130a, 051130b, 051205) show enhanced values of ozone, CO and CO2 between 355 and 380 K potential temperature in comparison with the first four flights (051116, 051119, 051123, 051125). Horizontal inmixing from the extra-tropical stratosphere and influence of the local convective system Hector cannot explain the enhanced values of the two flights on 30 November Therefore, other possible explanations for these enhanced CO, CO2 and ozone levels are proposed. The first explanation is vertical mixing in the vicinity of the jet stream. However, the jet cannot explain the differences between the flights on 30 November and the flights on 29 November and 5 December. Another possible explanation is influence of polluted boundary layer air masses from the Indonesian region. Especially air sampled during the flights on November 30 crossed large parts of northern Indonesia between 8 and 10 days before the measurements. Convective uplift of biomass burning and other pollution plumes can transport CO and ozone precursors into the upper troposphere, where they can significantly enhance the ozone production. The last chapter deals with the vertical ascent rate in the TTL and uses measurements of both the SCOUT-O3 and AMMA-SCOUT-O3 campaign as well as data from previous aircraft campaigns (TROCCINOX and APE-THESEO). Time scales and residence times for mean vertical transport in the background TTL are estimated for different seasons and over different geographic regions using in situ observations of CO2 and long-lived tracers. The vertical transport time scales are constrained using the seasonal variation of CO2 in the tropical troposphere as a “tracer clock” for vertical ascent. Two methods are applied to calculate the residence time in the layer between 360 and 390 K potential temperature. The first method uses the slope of the CO2 index, the second method fits the CO2 index directly to the measurements assuming a constant ascent rate. The first method yields residence times for Australia,West Africa, and Brazil of the same order, 35-45 days to 380 K and 50 days to 390 K (where no value can be derived for Australia as the slope is changing approximately one month before the campaign). For APE-THESEO, the method does not yield reasonable results. The best estimates using the second method show moderate residence times between 360 and 390 K of 60±25 days SCOUT-O3 (NH autumn) and 43±8 days for AMMA/SCOUT-O3 (NH summer). These results agree well with the results calculated using the first method. For APE-THESEO and TROCCINOX the best fits yield shorter residence times of 23±7 and 40±10 days, respectively, both during winter. These results correspond well to the expectations based on the seasonal variation of the Brewer-Dobson circulation.

Structured RNA regions are important gene control elements in prokaryotes and eukaryotes. Here, we show that the mRNA of a cyanobacterial heat shock gene contains a built-in thermosensor critical for photosynthetic activity under stress conditions. The exceptionally short 5´-untranslated region is comprised of a single hairpin with an internal asymmetric loop. It inhibits translation of the Synechocystis hsp17 transcript at normal growth conditions, permits translation initiation under stress conditions and shuts down Hsp17 production in the recovery phase. Point mutations that stabilized or destabilized the RNA structure deregulated reporter gene expression in vivo and ribosome binding in vitro. Introduction of such point mutations into the Synechocystis genome produced severe phenotypic defects. Reversible formation of the open and closed structure was beneficial for viability, integrity of the photosystem and oxygen evolution. Continuous production of Hsp17 was detrimental when the stress declined indicating that shutting-off heat shock protein production is an important, previously unrecognized function of RNA thermometers. We discovered a simple biosensor that strictly adjusts the cellular level of a molecular chaperone to the physiological need.
To date it is not clear at which stage of differentiation mature T cell leukaemia/lymphoma is initiated. Previous studies in our group showed that mature T cells are relatively resistant to transformation. We wanted to further investigate the transformation potential of NPM-ALK, p21SNFT and the viral oncoprotein Tax on mature T cells. First, we analyzed the effects on T cell growth in vitro after transducing human T cell lines with gammaretroviral vectors encoding these genes. No growth or proliferation promoting effect of all three genes was observed. In the second part of the project, we transduced murine, mature T cells and/or haematopoietic stem cells (HPCs/HSCs) and transplanted these cells into Rag-1 deficient recipients. All mice transplanted with NPM-ALK transduced monoclonal mature T cells (OT-1) developed leukaemia/lymphoma. In contrast, only few NPM-ALK transduced polyclonal T cell and HPC/HSC transplanted mice developed leukaemia/lymphoma. From the p21SNFT group, only two mice transplanted with transduced OT-1 T cells developed leukaemia/lymphoma, which showed high eGFP and interestingly CD19 expression. No malignancies were observed in Tax transplanted animals so far. Furthermore, the recipients do not show any eGFP marking in the periphery. In conclusion, our results show that compared to polyclonal T cells, monoclonal T cells are transformable after gammaretroviral transfer of NPM-ALK and p21SNFT.
Understanding causal relationships, or effective connectivity, between parts of the brain is of utmost importance because a large part of the brain’s activity is thought to be internally generated and, hence, quantifying stimulus response relationships alone does not fully describe brain dynamics. Past efforts to determine effective connectivity mostly relied on model based approaches such as Granger causality or dynamic causal modeling. Transfer entropy (TE) is an alternative measure of effective connectivity based on information theory. TE does not require a model of the interaction and is inherently non-linear. We investigated the applicability of TE as a metric in a test for effective connectivity to electrophysiological data based on simulations and magnetoencephalography (MEG) recordings in a simple motor task. In particular, we demonstrate that TE improved the detectability of effective connectivity for non-linear interactions, and for sensor level MEG signals where linear methods are hampered by signal-cross-talk due to volume conduction.
Sacoglossan sea slugs are unique in the animal kingdom in that they sequester and maintain active plastids that they acquire from the siphonaceous algae upon which they feed, making the animals photosynthetic. While most sacoglossan species digest their freshly ingested plastids within hours, four species from the family Plakobranchidae retain their stolen plastids (kleptoplasts) in a photosynthetically active state on time scales of weeks to months. The molecular basis of plastid maintenance within the cytosol of digestive gland cells in these photosynthetic metazoans is yet unknown, but is widely thought to involve gene transfer from the algal food source to the slugs based upon previous investigations of single genes. Indeed, normal plastid development requires hundreds of nuclear-encoded proteins, with protein turnover in photosystem II in particular known to be rapid under various conditions. Moreover, only algal plastids, not the algal nuclei, are sequestered by the animals during feeding. If algal nuclear genes are transferred to the animal either during feeding or in the germ line, and if they are expressed, then they should be readily detectable with deep-sequencing methods. We have sequenced expressed mRNAs from actively photosynthesizing, starved individuals of two photosynthetic sea slug species, Plakobranchus ocellatus Van Hasselt, 1824 and Elysia timida Risso, 1818. We find that nuclear-encoded, algal-derived genes specific to photosynthetic function are expressed neither in P. ocellatus nor in E. timida. Despite their dramatic plastid longevity, these photosynthetic sacoglossan slugs do not express genes acquired from algal nuclei in order to maintain plastid function.
Transcripts of NANOG and OCT4 have been recently identified in human t(4;11) leukemia and in a model system expressing both t(4;11) fusion proteins. Moreover, downstream target genes of NANOG/OCT4/SOX2 were shown to be transcriptionally activated. However, the NANOG1 gene belongs to a gene family, including a gene tandem duplication (named NANOG2 or NANOGP1) and several pseudogenes (NANOGP2-P11). Thus, it was unclear which of the NANOG family members were transcribed in t(4;11) leukemia cells. 5'-RACE experiments revealed novel 5'-exons of NANOG1 and NANOG2, which could give rise to the expression of two different NANOG1 and three different NANOG2 protein variants. Moreover, a novel PCR-based method was established that allows distinguishing between transcripts deriving from NANOG1, NANOG2 and all other NANOG pseudogenes (P2–P11). By applying this method, we were able to demonstrate that human hematopoietic stem cells and different leukemic cells transcribe NANOG2. Furthermore, we functionally tested NANOG1 and NANOG2 protein variants by recombinant expression in 293 cells. These studies revealed that NANOG1 and NANOG2 protein variants are functionally equivalent and activate a regulatory circuit that activates specific stem cell genes. Therefore, we pose the hypothesis that the transcriptional activation of NANOG2 represents a ‘gain-of-stem cell function’ in acute leukemia.
Nusa Tenggara Timor, a south-eastern province of Indonesia, is populated mainly by Christians. The Alor-Pantar Archipelago has a majority of Protestant inhabitants who were baptized by Dutch Calvinists in the first half of the twentieth century. In addition, there are some coastal enclaves that have been inhabited by Muslims for centuries. In some areas, such as in the headland of Muna (Tanjung Muna) forming the northeast of Pantar Island, there is an even greater diversity of monotheistic religions, with some Catholic families living next to Protestants and Muslims. All adherers of the three religious faiths living at Tanjung Muna share core elements of the local adat, which consists of core rules relating to social behavior. It is believed that the ancestors will notice transgressions of these rules, and may use their supernatural power to punish their human descendants. In Indonesia, the term adat was first used by Muslims to distinguish the non-Islamic practices from Muslim faith (Keane 1997:260-261). This is definitely not the case in the village of Pandai at the coast of Tanjung Muna, where Islam tolerates ancestral worship. The same is true for the Catholics in the inland village of Helangdohi, who do not only tolerate but even support such customs. Some villagers from Helangdohi had become acquainted with this kind of Catholicism on the nearby island of Flores, where ancestral worship is encouraged by the missionaries of the Societas Verbi Divini (SVD). The attitude of Protestantism, at least in the Alor Archipelago, is quite the contrary of the permissive views held by Catholicism and Islam. In the 1930s the Protestant-Calvinist missionaries banned any kind of ancestral worship and destroyed most relics (Dalen 1928: Picture 1). These drastic measures demanded the disavowal of the ancestors, including the destruction of heirlooms and omitting of rituals.
This paper investigates the accuracy and heterogeneity of output growth and inflation forecasts during the current and the four preceding NBER-dated U.S. recessions. We generate forecasts from six different models of the U.S. economy and compare them to professional forecasts from the Federal Reserve’s Greenbook and the Survey of Professional Forecasters (SPF). The model parameters and model forecasts are derived from historical data vintages so as to ensure comparability to historical forecasts by professionals. The mean model forecast comes surprisingly close to the mean SPF and Greenbook forecasts in terms of accuracy even though the models only make use of a small number of data series. Model forecasts compare particularly well to professional forecasts at a horizon of three to four quarters and during recoveries. The extent of forecast heterogeneity is similar for model and professional forecasts but varies substantially over time. Thus, forecast heterogeneity constitutes a potentially important source of economic fluctuations. While the particular reasons for diversity in professional forecasts are not observable, the diversity in model forecasts can be traced to different modeling assumptions, information sets and parameter estimates. Tracer measurements in the tropical tropopause layer during the AMMA/SCOUT-O3 aircraft campaign

We present airborne in situ measurements made during the AMMA (African Monsoon Multidisciplinary Analysis)/SCOUT-O3 campaign between 31 July and 17 August 2006 on board the M55 Geophysica aircraft, based in Ouagadougou, Burkina Faso. CO<sub>2</sub> and N<sub>2</sub>O were measured with the High Altitude Gas Analyzer (HAGAR), CO was measured with the Cryogenically Operated Laser Diode (COLD) instrument, and O<sub>3</sub> with the Fast Ozone ANalyzer (FOZAN). We analyze the data obtained during five local flights to study the dominant transport processes controlling the tropical tropopause layer (TTL) above West-Africa: deep convection up to the level of main convective outflow, overshooting of deep convection, horizontal inmixing across the subtropical tropopause, and horizontal transport across the subtropical barrier. Except for the flight of 13 August, distinct minima in CO<sub>2</sub> indicate convective outflow of boundary layer air in the TTL. The CO<sub>2</sub> profiles show that the level of main convective outflow was mostly located between 350 and 360 K, and for 11 August reached up to 370 K. While the CO<sub>2</sub> minima indicate quite significant convective influence, the O<sub>3</sub> profiles suggest that the observed convective signatures were mostly not fresh, but of older origin. When compared with the mean O<sub>3</sub> profile measured during a previous campaign over Darwin in November 2005, the O<sub>3</sub> minimum at the main convective outflow level was less pronounced over Ouagadougou. Furthermore O<sub>3</sub> mixing ratios were much higher throughout the whole TTL and, unlike over Darwin, rarely showed low values observed in the regional boundary layer. Signatures of irreversible mixing following overshooting of convective air were scarce in the tracer data. Some small signatures indicative of this process were found in CO<sub>2</sub> profiles between 390 and 410 K during the flights of 4 and 8 August, and in CO data at 410 K on 7 August. However, the absence of expected corresponding signatures in other tracer data makes this evidence inconclusive, and overall there is little indication from the observations that overshooting convection has a profound impact on TTL composition during AMMA. We find the amount of photochemically aged air isentropically mixed into the TTL across the subtropical tropopause to be not significant. Using the N<sub>2</sub>O observations we estimate the fraction of aged extratropical stratospheric air in the TTL to be 0.0±0.1 up to 370 K during the local flights, increasing above this level to 0.2±0.15 at 390 K. The subtropical barrier, as indicated by the slope of the correlation between N<sub>2</sub>O and O<sub>3</sub> between 415 and 490 K, does not appear as a sharp border between the tropics and extratropics, but rather as a gradual transition region between 10 and 25° N latitude where isentropic mixing between these two regions may occur.
As an exemplum of that kind of “modern” art, in terms of Adorno, Kafka’s work is marked not only by its strictly “realistic” character, but also by the unavoidable critical and testimonial value of that realism. According to this perspective, both in Adorno and in Benjamin the testimonial aspect of Kafkian writing – that is of a writing as “dialectical image”, as memory of the unfullfilled possibility – it’s all the same not with its symbolical or “epiphanical” aspect but instead with its “allegorical” one.
A lot of effort in lattice simulations over the last years has been devoted to studies of the QCD deconfinement transition. Most state-of-the-art simulations use rooted staggered fermions, while Wilson fermions are affected by large systematic uncertainties, such as coarse lattices or heavy sea quarks. Here we report on an ongoing study of the transition, using two degenerate flavours of nonperturbatively O(a) improved Wilson fermions. We start with Nt = 12 and 16 lattices and pion masses of 600 to 450 MeV, aiming at chiral and continuum limits with light quarks.
Orientation: The work of industrial/organisational (I/O) psychologists presents an interesting and relevant context for studying meaning and engagement as components of happiness. Research purpose: The aim of this study was to determine how I/O psychologists experience the meaning of their work and to investigate the relationships between their experiences of work-role fit, meaning of work, psychological meaningfulness and work engagement, utilising the happiness framework proposed by Seligman (2002). Motivation for the study: I/O psychologists spend more than 88% of their working day with people, and they are primary role models for happiness in the workplace. Information about their work engagement and experiences of meaning is therefore needed. Research design, approach and method: A survey design was used. A convenience sample (n = 106) was taken of I/O psychologists in South Africa. A biographical questionnaire, the Work-Role Fit Scale, the Work-Life Questionnaire, the Psychological Meaningfulness Scale, the Work Engagement Scale and a survey measuring the actual and desired time spent on six broad categories of work were administered. Main findings: Work-role fit predicted psychological meaningfulness and work engagement. The calling orientation to work predicted both psychological meaningfulness and work engagement. Work-role fit mediated the relationship between the meaning of work and psychological meaningfulness. Work-role fit partially mediated the relationship between a calling orientation to work and work engagement. Practical implications: A calling orientation to work should be fostered in I/O psychologists because it contributes to experiences of work-role fit, psychological meaningfulness and work engagement. Contribution/value-add: The results of this study contribute to scientific knowledge about work-role fit, engagement and meaning as components of happiness of I/O psychologists.
Towards correctness of program transformations through unification and critical pair computation

Correctness of program transformations in extended lambda-calculi with a contextual semantics is usually based on reasoning about the operational semantics which is a rewrite semantics. A successful approach is the combination of a context lemma with the computation of overlaps between program transformations and the reduction rules, which results in so-called complete sets of diagrams. The method is similar to the computation of critical pairs for the completion of term rewriting systems. We explore cases where the computation of these overlaps can be done in a first order way by variants of critical pair computation that use unification algorithms. As a case study of an application we describe a finitary and decidable unification algorithm for the combination of the equational theory of left-commutativity modelling multi-sets, context variables and many-sorted unification. Sets of equations are restricted to be almost linear, i.e. every variable and context variable occurs at most once, where we allow one exception: variables of a sort without ground terms may occur several times. Every context variable must have an argument-sort in the free part of the signature. We also extend the unification algorithm by the treatment of binding-chains in let- and letrec-environments and by context-classes. This results in a unification algorithm that can be applied to all overlaps of normal-order reductions and transformations in an extended lambda calculus with letrec that we use as a case study.
Introduction: It has been proposed that individual genetic variation contributes to the course of severe infections and sepsis. Recent studies of single nucleotide polymorphisms (SNPs) within the endotoxin receptor and its signaling system showed an association with the risk of disease development. This study aims to examine the response associated with genetic variations of TLR4, the receptor for bacterial LPS, and a central intracellular signal transducer (TIRAP/Mal) on cytokine release and for susceptibility and course of severe hospital acquired infections in distinct patient populations. Methods: Three intensive care units in tertiary care university hospitals in Greece and Germany participated. 375 and 415 postoperative patients and 159 patients with ventilator associated pneumonia (VAP) were included. TLR4 and TIRAP/Mal polymorphisms in 375 general surgical patients were associated with risk of infection, clinical course and outcome. In two prospective studies, 415 patients following cardiac surgery and 159 patients with newly diagnosed VAP predominantly caused by Gram-negative bacteria were studied for cytokine levels in-vivo and after ex-vivo monocyte stimulation and clinical course. Results: Patients simultaneously carrying polymorphisms in TIRAP/Mal and TLR4 and patients homozygous for the TIRAP/Mal SNP had a significantly higher risk of severe infections after surgery (odds ratio (OR) 5.5; confidence interval (CI): 1.34 - 22.64; P = 0.02 and OR: 7.3; CI: 1.89 - 28.50; P < 0.01 respectively). Additionally we found significantly lower circulating cytokine levels in double-mutant individuals with ventilator associated pneumonia and reduced cytokine production in an ex-vivo monocyte stimulation assay, but this difference was not apparent in TIRAP/Mal-homozygous patients. In cardiac surgery patients without infection, the cytokine release profiles were not changed when comparing different genotypes. Conclusions: Carriers of mutations in sequential components of the TLR signaling system may have an increased risk for severe infections. Patients with this genotype showed a decrease in cytokine release when infected which was not apparent in patients with sterile inflammation following cardiac surgery.
Iron uptake is an essential process in all Gram-negative bacteria including cyanobacteria and therefore different transport systems evolved during evolution. In cyanobacteria, however, the iron demand is higher than in proteobacteria due to the function of iron as cofactor in e.g. photosynthesis and nitrogen fixation. Most of the transport systems depend on outer membrane localized TonB-dependent transporters (TBDTs), a periplasma-facing TonB protein and a plasma membrane localized machinery (ExbBD). So far, iron chelators (siderophores), oligosaccharides and polypeptides have been identified as substrates of TBDTs. However, in proteobacteria TonB-dependent outer membrane transporter represent a well-explored subject whereas for cyanobacteria almost nothing is known about possible TonB-dependent uptake systems for iron or other substrates. The heterocyst-forming filamentous cyanobacterium Anabaena sp. PCC 7120 is known to secrete the siderophore schizokinen, but its transport system has remained unidentified. For Anabaena sp. PCC 7120 22 genes were identified as putative TBDTs covering almost all known TBDT subclasses. This is a high number of TBDTs compared to other cyanobacteria. The expression of the 22 putative TBDTs individually depends on the presence of iron, copper or nitrogen. The atypical dependence of TBDT gene expression on different nutrition points to a yet unknown regulatory mechanism. In addition, the hypothesis of the absence of TonB in Anabaena sp. PCC 7120 was clarified by the identification of an according sequence, all5036. Inspection of the genome of Anabaena sp. PCC 7120 shows that only one gene encoding a putative TonB-dependent iron transporter, namely alr0397, is positioned close to genes encoding enzymes involved in the biosynthesis of a hydroxamate siderophore. The expression of alr0397 was elevated under iron-limited conditions. Inactivation of this gene caused a moderate phenotype of iron starvation in the mutant cells. The characterization of the mutant strain showed that Alr0397 is a TonB-dependent schizokinen transporter (SchT) of the outer membrane and that alr0397 expression and schizokinen production are regulated by the iron homeostasis of the cell. Additional two genes of Anabaena sp. PCC 7120 involved in this process were identified. SchE encoded by all4025 is a putative cytoplasmic membrane-localized transporter involved in TolC-dependent siderophore secretion. The mutation of schE resulted in an enhanced sensitivity to high metal concentrations and in drastically reduction of secretion of hydroxamate-type siderophores. IacT coded by all4026 is a predicted outer membrane-localized TonB-dependent iron transporter. Inactivation of iacT resulted in reduced sensitivity to elevated iron and copper levels, whereas decoupling the expression from putative regulation by exchange of the promoter resulted in sensitization against tested metals. Further analysis showed that iron and copper effects are synergistic because decrease of iron induced a significant decrease of copper levels in the iacT insertion mutant but an increase of those levels in Anabaena sp. PCC 7120 where expression of all4026 is under the trc-promoter. In consequence, the results unravel a link between iron and copper homeostasis.
Leukotrienes (LTs) are pro-inflammatory lipid mediators that belong to the group of eicosanoids, which are oxygenated metabolites of one common precursor, the aracidonic acid (AA). This polyunsaturated fatty acid is esterified at the sn-2 position of cellular membrane phospholipids and can be released by cytosolic phospholipase A2 alpha (cPLA2alpha) enzymatic deacylation. AA can be converted into LTs by the catalytic reaction of 5-lipoxygenase (5-LO). Enzymatic activation of cPLA2alpha as well as of 5-LO is regulated by similar determinants. In response to cellular stimuli that elevate the intracellular Ca2+ level and/or activate MAP kinase pathways, cPLA2alpha and 5-LO comigrate from a soluble cell compartment (mainly the cytosol) to the nuclear membrane, where AA is released und converted into LTs. LTs play a significant role in promoting inflammatory reactions and immune processes. They have been shown to be released from leukocytes in response to bacterial and viral infections and substantially contribute to an effective immune reaction for host defense. Innate immune pathogen recognition is mediated to a substantial part by the Toll-like receptor (TLR) family. So far, 10 human TLR subtypes have been identified, all of which detect distinct highly conserved microbial structures and trigger the induction of signaling pathways that lead to the expression of numerous immune and inflammatory genes. TLR signaling culminates in the activation NF-kappaB and/or MAP kinases, which as well are known to be involved in the regulation of cellular LT biosynthesis. In this regard, it seemed conceivable that the release of LTs might be regulated by TLR activation. Present studies were undertaken in order to verify and characterize a possible influence of TLR activation on the LT biosynthesis, and furthermore to identify the involved signaling pathways and underlying mechanisms. First experiments revealed that pre-incubation of differentiated Mono Mac 6 (MM6) cells with a TLR4 ligand, a TLR5 ligand, as well as with different TLR2 ligands led to an about 2-fold enhancement of Ca2+ ionophore induced LT biosynthesis. Ligands of other TLR subtypes did not show any influence. These observations could also be confirmed in primary human monocytes stimulated with ionophore or fMLP. With focus on TLR2 ligands, further studies were carried out to characterize the observed enhancement of LT biosynthesis in MM6 cells. It was demonstrated that the extent of LT formation was dependent on the ligand concentration used, but was also dependent on the duration of pre-incubation. Ligand pre-incubation of 15 minutes was optimal to maximally enhance LT formation and further prolongation of pre-incubation decreased LT formation again. Moreover, simultaneous addition of TLR2 ligands with ionophore did also not enhance LT formation. These results indicated that TLR2 ligands seemed to prime human monocytes for an enhanced response upon ionophore stimulation, but did not act as costimuli, which per se were not capable of directly stimulating the biosynthesis of LTs. To analyze the underlying mechanism, the impact of TLR2 ligands on the two key enzymes of the LT biosynthesis pathway, cPLA2alpha and 5-LO, was investigated. In this regard, 5-LO could not been shown to be positively regulated by TLR ligand priming. Neither a direct stimulation, nor an enhancement of 5-LO activity by TLR ligands was detectable in MM6 cells. Similarly, TLR2 ligands did also not enhance ionophore induced 5-LO translocation to the nuclear membrane. However, it was shown that TLR2 ligands enhanced ionophore induced release of AA in MM6 cells, which occurred with a similar time course as LT formation, displaying a maximum at 10 minutes of pre-incubation. A direct stimulation of AA release, however, could not been detected. Inhibitor studies revealed cPLA2alpha to be essential for AA release in TLR2 ligand primed, ionophore stimulated MM6 cells, but also sPLA2 was found to be involved. However, the priming effect of TLR2 ligands was mediated exclusively by cPLA2alpha. Western Blot analyses revealed that p38 MAP kinase, as well as ERK1/2, are activated in MM6 cells in response to TLR2 ligands, and also Ser-505 phosphorylation of cPLA2alpha was detected, which is known to be mediated by MAP kinases and to increase cPLA2alpha activity in vitro. Maximal cPLA2alpha phosphorylation occurred after 5-10 minutes of TLR2 ligand incubation, slightly preceding maximal AA release at 10 minutes and maximal LT formation at 15 minutes of priming. The combined use of a specific p38 MAPK inhibitor with an inhibitor of the ERK1/2 signaling pathway resulted in a complete prevention of cPLA2alpha phosphorylation and TLR2 ligand mediated enhancement of AA release. Thus, both MAPK pathways seem to play a role for TLR2 ligand mediated priming effects on the release of AA. An impact of other kinases such as Mnk-1 and CamKII, which can also regulate cPLA2alpha by phosphorylation, was excluded. Finally, an anti-hTLR2 antibody significantly reduced enhanced AA release, confirming the priming effects to be dependent on TLR2 activation. In summary, it was concluded that the increase of LT biosynthesis by TLR2 ligand priming is considerably due to an enhanced cellular AA supply, which arises from a MAPK mediated phosphorylation and up-regulation of cPLA2alpha. TLR dependent enhancement of LT biosynthesis represents an interesting link between activation of innate immune receptors and the rapid formation of pro-inflammatory lipid mediators. On the one hand, this support the role of LTs in host defence and infectious diseases, but may also be relevant in pathophysiological processes, which involve TLRs as well as LTs, as it has been shown for the pathogenesis of atherosclerosis or allergic diseases.
Suicide genes have been broadly used in gene therapy. They can serve as safety tools for conditional elimination of infused cells or for directed tumor therapy. To date, the Herpes simplex virus thymidine kinase/ ganciclovir (HSVtk/GCV) system is the most prominent and the most widely used suicidegene/prodrug combination. Despite its promising performance, the system displays limitations, which include relatively slow killing kinetics and toxicity of the prodrug GCV. Consequently, several groups have either developed new suicide-gene/prodrug combinations or attempted to improve the established HSVtk/GCV suicide system. The present study also aimed towards optimization of the HSVtk/GCV system. To do so, a novel, codon-optimized point mutant (A168H) of HSVtk was developed. The novel mutant was named TK.007. It was extensively tested for its efficiency in two relevant settings: (1) control of severe graft-versus-host disease (GvHD) after adoptive immunotherapy with Tlymphocytes, and (2) direct elimination of targeted tumor cells. TK.007 was compared to the broadly used wild-type, splice-corrected scHSVtk and to a codon-optimized HSVtk (coHSVtk) not bearing the above point mutation. (1) For experiments related to the adoptive immunotherapy approach, HSVtkvariants were expressed from a γ-retroviral MP71 vector as a fusion construct with the selection and marker gene tCD34. Expression levels for TK.007 in transduced lymphoid and myeloid cell lines were significantly higher at initial transduction and over a 12 week period compared to the commonly used scHSVtk and coHSVtk indicating reduced toxicity of TK.007. Killing kinetics of transduced cell lines (PM1 and K562) and primary human T cells were significantly faster for TK.007 in comparison to scHSVtk and coHSVtk in vitro. In vivo-functionality of TK.007 was assessed in an allogeneic transplantation model. T cells derived from C57BL/6J.Ly5.1 donor mice were transduced with MP71 vectors expressing scHSVtk or TK.007. Transduced cells were selected and transplanted into Balb/c Rag2-/- γ-/- immune-deficient recipient mice. Acute, severe GvHD occurred and was effectively abrogated in all mice transplanted with TK.007- transduced T cells, and in five out of six mice transplanted with scHSVtk-transduced cells. In a slightly modified quantitative allogeneic transplantation mouse model, significantly faster and more efficient in vivo killing was demonstrated for TK.007 as compared to scHSVtk, especially at low doses of GCV. (2) In order to assess TK.007 functionality in cells derived from solid tumors, HSVtk-variants were expressed from lentiviral gene ontology (LeGO) vectors in combination with an eGFP/neo-opt selection cassette. Transduced and selected tumor cell lines that derived from several tissues were eliminated at significantly lower GCV doses and to higher extents when transduced with TK.007 compared to scHSVtk. Moreover, a significantly stronger bystander effect of TK.007 was demonstrated. The superior in vitro efficiency of TK.007 was confirmed in an in vivo subcutaneous xenograft mouse model for glioblastoma in NOD/SCID mice. Mice transplanted with TK.007 transduced cells stayed tumor-free after treatment with different GCV-doses. On the contrary, mice of the scHSVtk group either demonstrated only transiently reduced tumor growth in the low-dose GCV group (10 mg/kg) compared to the control groups or suffered from relatively fast relapses after initial tumor shrinking in the standarddose (50 mg/kg) GCV group. As a result, all mice in the scHSVtk group died from vigorous tumor growth. In summary, in two different applications for suicide gene therapy the present study has demonstrated superior functional performance of the novel suicide gene TK.007 as compared to the broadly used wild-type scHSVtk. Differences became particularly pronounced at low doses of GCV. It can be concluded that the new TK.007-gene represents a promising alternative to the commonly used scHSVtk for gene therapeutic applications.
Background: Thymostimulin is a thymic peptide fraction with immune-mediated cytotoxicity against hepatocellular carcinoma (HCC) in vitro and palliative efficacy in advanced HCC in two independent phase II trials. The aim of this study was to assess the efficacy of thymostimulin in a phase III trial. Methods: The study was designed as a prospective randomised, placebo-controlled, double-blind, multicenter clinical phase III trial. Between 10/2002 and 03/2005, 135 patients with locally advanced or metastasised HCC (Karnofsky [greater than or equal to]60% / Child-Pugh [less than or equal to]12) were randomised to receive thymostimulin 75 mg s.c. 5x/week or placebo stratified according to liver function. Primary endpoint was twelve-month survival, secondary endpoints overall survival (OS), time to progression (TTP), tumor response, safety and quality of life. A subgroup analysis according to liver function, KPS and tumor stage (Okuda, CLIP and BCLC) formed part of the protocol. Current Controlled Trials ISRCTN64487365. Results: Twelve-month survival was 28% [95%CI 17-41; treatment] and 32% [95%CI 19-44; control] with no significant differences in median OS (5.0 [95% CI 3.7-6.3] vs. 5.2 [95% CI 3.5-6.9] months; p=0.87, HR=1.04 [95% CI 0.7-1.6]) or TTP (5.3 [95%CI 2.0-8.6] vs. 2.9 [95%CI 2.6-3.1] months; p=0.60, HR=1.13 [95% CI 0.7-1.8]). Adjustment for liver function, Karnofsky status or tumor stage did not affect results. While quality of life was similar in both groups, fewer patients on thymostimulin suffered from accumulating ascites and renal failure. Conclusions: In our phase III trial, we found no evidence of any benefit to thymostimulin in the treatment of advanced HCC and there is therefore no justification for its use as single-agent treatment. The effect of thymostimulin on hepato-renal function requires further confirmation. trial registration: Current Controlled Trials ISRCTN64487365

Products of ene reactions of isobutene and tBu2Si=N-SiRtBu2 (R = tBu, Ph) are formed. X-Ray quality crystals of H2C=C(CH2SitBu2-NH-SiPhtBu2)2 (monoclinic, space group C2/c, Z = 4) were grown from a benzene solution at ambient temperature, whereas single crystals of H2C=C(CH2SitBu2-NH-SitBu3)2 (monoclinic, space group P21, Z = 2) were obtained by recrystallization from THF.
Therapy of hemorrhagic shock with following resuscitation-induced liver injury : in vivo study

Shock resulting from life-threatening blood-loss (hemorrhagic shock) represents the most frequent injury pattern after a traumatic insult. Hemorrhagic shock induces inflammatory changes, characterized by highly complex pathophysiological pathways often resulting in death. In this study, we establish an experimental in vivo model of H/R in rats and study the mechanisms which determine the hepatic injury after H/R. Furthermore, we show that hemorrhagic shock with following resuscitation is accompanied with release of systemic and local pro-inflammatory mediators, increased infiltration of hepatic neutrophils in the liver, increased oxidative and nitrosative stress, enhanced cell death of both types, apoptosis and necrosis, conspicuous cytoskeletal rearrangements, loss of hepatic integrity and finally high general mortality rates, up to 80%. In addition, the effects of two potential therapeutic interventions to prevent the H/R induced liver injury are explored in a model of H/R in rats. First, the role of JNK and its inhibition by D-JNKI-1 in preservation of hepatic integrity following H/R was analyzed. Second, we investigated the potential of simvastatin to prevent the disturbed inflammatory response and hepatic injury after H/R. The effects of both therapeutic interventions were studied by looking at several inflammatory parameters, markers of oxidative and nitrosative stress, cytoskeleton integrity, microcirculatory parameters, underlying signaling cascades, liver damage and mortality. Highly specific blockade of JNK with the potent, inhibitory peptide D-JNKI-1 revealed the crucial role of the JNK signaling pathway in the H/R induced pathophysiology and strong protective effects of DJNKI- 1 in H/R induced liver injury, when the peptide was applied before and even after hemorrhagic shock. The other therapeutic intervention tested in this study was the use of simvastatin which also revealed protective effects after H/R and even a remarkable improvement in survival after H/R. We show that H/R induced release of pro-inflammatory cytokines, hepatic PMNL infiltration, increased oxidative and nitrosative stress, apoptosis and necrosis can be diminished by treatment with D-JNKI-1 but also with simvastatin in vivo. Furthermore, simvastatin reduces H/R induced cytoskelatal rearrangements, loss of liver integrity and the mortality rate after H/R. The key pathway which underlies these beneficial effects of simvastatin is the Rho kinase pathway. Identification of both mechanisms as well as the effectiveness of both substances provide new insights in the close interaction between hypoxia and the immune system and present a promising basis for the anti-inflammatory, hepatoprotective treatment after H/R.
Das Ziel dieser retrospektiven Untersuchung ist, die medikamentöse Therapie des neonatalen Entzugsyndroms mittels Clonidin und Chloralhydrat mit der sehr häufig angewendeten Kombinationstherapie aus Morphin und Phenobarbital zu vergleichen. Im dem Zeitraum zwischen 1998 und 2008 wurden in unserer Klinik 133 Neugeborene mit einem neonatalen Entzugssyndrom behandelt. Bei allen Patienten handelte es sich um Neugeborene drogenabhängiger Mütter, welche während der Schwangerschaft mit Methadon substituiert wurden. Bei 29 Patienten wurde eine Therapie mit Clonidin und Chloralhydrat und bei 64 Patienten eine Entzugstherapie mittels Morphin und Phenobarbital durchgeführt. In der Clonidingruppe konnte eine signifikant kürzere Behandlungsdauer (Median: 14 d vs. 35 d) festgestellt werden. Entsprechend war auch die gesamte Aufenthaltsdauer in der Clonidingruppe deutlich kürzer (Median: 32 d vs. 44 d). Außerdem konnte in der Clonidingruppe ein deutlich milderer Verlauf der Entzugssymptomatik festgestellt werden.
In the present work, the problem of protein folding is addressed from the point of view of equilibrium thermodynamics. The conformation of a globular protein in solution at common temperatures is quite complicated without any geometrical symmetry, but it is an ordered state in the sense of its biological activity. This complicated conformation of a single protein molecule is destroyed upon increasing the temperature or by the addition of appropriate chemical agents, as is revealed by the loss of its activity and change of the physical properties, and so on. Once the complicated native structures having biological activity are lost, it would be natural to suppose that the native structure could hardly be restored. Nevertheless, pioneers, such as Anson and Mirsky, recognized as early as in 1925 that this was not always the case. If one defines the folded and unfolded states of a protein as two distinct phases of a system, then under the variation of temperature the system is transformed from one phase state into another and vice versa. The process of protein folding is accompanied by the release or absorption of a certain amount of energy, corresponding to the first-oder-type phase transitions in the bulk. Knowing the partition function of the system one can evaluate its energy and heat capacity under different temperatures. This task was performed in this work. The results of the developed statistical mechanics model were compared with the results of molecular dynamic simulations of alanine poylpeptides. In particular, the dependencies on temperature of the total energy of the system and heat capacity were compared for alanine polypeptides consisting of 21, 30, 40, 50 and 100 amino acids. The good correspondence of the results of the theoretical model with the results of molecular dynamics simulations allowed to validate the assumptions made about the system and to establish the accuracy range of the theory. In order to perform the comparison of the results of theoretical model and the molecular dynamics simulations it is necessary to perform the efficient analysis of the results of molecular dynamics simulations. This task was also addressed in the present work. In particular, different ways to obtain dependence of the heat capacity on temperature from molecular dynamics simulations are discussed and the most efficient one is proposed. The present thesis reports the result of molecular dynamic simulations for not only alanine polypeptides by also for valine and leucine polypeptides. In valine and leucine polypeptides, it is also possible to observe the helix↔random coil transitions with the increase of temperature. The current thesis presents a work that starts with the investigation of the fundamental degrees of freedom in polypeptides that are responsible for the conformational transitions. Then this knowledge is applied for the statistical mechanics description of helix↔coil transitions in polypeptides. Finally, the theoretical formalism is generalized for the case of proteins in water environment and the comparison of the results of the statistical mechanics model with the experimental measurements of the heat capacity on temperature dependencies for two globular proteins is performed. The presented formalism is based on fundamental physical properties of the system and provides the possibility to describe the folding↔unfolding transitions quantitatively. The combination of these two facts is the major novelty of the presented approach in comparison to the existing ones. The “transparent” physical nature of the formalism provides a possibility to further apply it to a large variety of systems and processes. For instance, it can be used for investigation of the influence of the mutations in the proteins on their stability. This task is of primary importance for design of novel proteins and drug delivering molecules in medicine. It can provide further insights into the problem of protein aggregation and formation of amyloids. The problem of protein aggregation is closely associated with various illnesses such as Alzheimer and mad cow disease. With certain modifications, the presented theoretical method can be applied to the description of the protein crystallization process, which is important for the determination of the structure of proteins with X-Rays. There many other possible applications of the ideas described in the thesis. For instance, the similar formalism can be developed for the description of melting and unzipping of DNA, growth of nanotubes, formation of fullerenes, etc.
This paper argues that the Fairtrade certification system represents an illuminating example of the challenge of systematically determining consumer and entrepreneurial responsibilities in our global age. In taking up the central question of what, if anything, may be called ‘just’ or ‘fair’ in Fairtrade, I more precisely argue for a two-fold thesis: that (1) a meaningful evaluation of Fairtrade must consider both an interactional and an (arguably prior) institutional understanding of global responsibilities to promote justice and that (2) Fairtrade can be better defended against several popular objections from the perspective of a theory that adequately differentiates between interactional responsibilities and institutional responsibilities of promoting justice under unjust circumstances.
AS A MATTER OF COURSE, QUANTITATIVE DATA SUCH AS TIME SERIES AND QUARTERLY FIGURES ARE FREQUENTLY USED IN FINANCIAL ANALYSIS. SUCH DATA CAN BE PROCESSED AUTOMATICALLY AND INTERPRETED RATHER EFFICIENTLY. HOWEVER, A SIGNIFICANT PERCENTAGE OF RELEVANT INFORMATION ORIGINATES FROM UNSTRUCTURED SOURCES, PRIMARILY TEXTUAL DATA, WHICH REQUIRE MANUAL (HUMAN) INTERPRETATION. WE EXPLORE EMPIRICALLY HOW MACHINE LEARNING TECHNIQUES CAN PROVIDE SUPPORT FOR ANALYZING AND INTERPRETING SUCH TEXTUAL DATA SOURCES.
The concept of embeddedness plays a central role in the segment of economic sociology and social theory which is inspired by the works of Karl Polanyi. But to the extent that embeddedness is understood in a substantialist manner, implying the existence of a unitary lifeworld, the desire for embeddedness is an impossible aspiration under modern conditions. Throughout the modern era it is however possible to observe the emergence of complex societal stabilization mechanisms, which serve as substitutes to traditional forms of embeddedness. The emergence of function specific cultures, in the form of, for example, legal, political and scientific cultures, establishing a ‘second nature’ in the Hegelian sense, is one example of this. Other examples are (neo-)corporatist institutions which fulfilled a central stabilising role in classical modernity and the kind of network based governance arrangements which fulfil a similar position in today’s radicalised modernity.
We compute the static-light baryon spectrum with Nf = 2 flavors of sea quarks using Wilson twisted mass lattice QCD. As light valence quarks we consider quarks, which have the same mass as the sea quarks with corresponding pion masses in the range 340MeV<∼ mPS<∼ 525MeV, as well as partially quenched quarks, which have the mass of the physical s quark. We extract masses of states with isospin I = 0,1/2,1, with strangeness S = 0,−1,−2, with angular momentum of the light degrees of freedom j = 0,1 and with parity P = +,−. We present a preliminary extrapolation in the light u/d and an interpolation in the heavy b quark mass to the physical point and compare with available experimental results.
Tumor hypoxia and nutrient starvation are common phenomena in cancerous tissue. Cells that resist this hostile environment are selected for a more aggressive phenotype, usually accompanied by therapy resistance. The hypoxia inducible factors HIF-1a and HIF-2a play a key role in the adaptive homeostatic responses to these challenging conditions inducing a number of target genes that are involved in the regulation of a variety of cellular processes such as angiogenesis, proliferation, metabolism, self-renewal and cell death/cycle arrest. Thus, the HIF pathway encompasses opposing adaptive responses on tumor growthgrowth promoting abilities on the one hand and growth inhibiting on the other. A recent study in our lab uncovered that this switch between cell death and cell survival critically depends on HIF-2a protein levels. Since PHDs (HIF prolyl hydroxylases) are the main regulators of HIF protein abundance and hypoxia drives the malignant phenotype of tumors, we wanted to characterize HIF regulatory functions of PHDs under hypoxic conditions. Our intention was to reveal the importance of PHD contribution to the opposing functions of HIFs under hypoxia. Characterization of PHD1-4 mRNA and protein expression levels under normoxic and hypoxic conditions in glioblastoma cell lines led to the identification of PHD2 and PHD3 as hypoxia inducible PHD isoforms and highlighted their predominant function under hypoxia. Mechanistically, we demonstrated that HIF mediates the hypoxic induction of PHD2 and 3 within a negative feedback loop, promoting its own degradation during prolonged hypoxia. The functional impact of PHD2 and 3 abundance on cell viability under hypoxic conditions was analyzed by disrupting PHD2 and PHD3 function either through a siRNA mediated approach or by application of the PHD inhibitor DMOG. These experiments uncovered that PHD2 and 3 are protective under hypoxic conditions and that PHD inhibition expedites cell death. Combined HIF and PHD suppression under hypoxic conditions abrogated this increased susceptibility to cell death, clearly showing that PHD2 and 3 act in a negative feedback regulatory loop to limit the HIF response under prolonged hypoxia. With respect to possible future therapeutical applications we co-treated cells with a PHD inhibitor and pro-apoptotic agents staurosporine or TRAIL. Co-challenging tumor cells even potentiated the cell death response, indicating a more widespread protective function of PHD. Taken together PHD2 and 3 protect tumor cells from cell death induction, functioning in a negative feedback regulatory loop to constrain the HIF dependent cell death responses under hypoxia. Interestingly, however, when assessing the role of PHD2 and PHD3 in in vivo tumor growth using an intracranial tumor model, we identified an exclusive tumor suppressor function for PHD3. Loss of PHD3 function enhanced tumor growth whereas increased PHD3 expression diminished the tumor burden. The accelerated tumor growth following PHD3 loss could be attributed to a decrease in the induction of apoptosis and an increase in proliferation. Tumor cells are frequently exposed to temporary and spatial depletion of nutrients. Interestingly, PHD3 loss conferred a growth advantage under growth factor deprivation. The growth regulatory function of PHD3 was isoform specific, HIF independent and importantly, did not require the hydroxylase function of PHD3. Previous reports have uncovered a regulatory function of the PHD system in NF-kB signaling. However, our results demonstrated that NF- kB signaling remained unaffected by alteration in the PHD3 status of the cell. Additionally, the PHD3 tumor suppressor function proved to be independent of two putative PHD3 downstream effectors, ATF4 and KIF1Bb. Mechanistically, PHD3 suppression reduced EGFR internalization, enhancing the amount of EGFR expressed on the cell surface. We further showed that the impaired EGFR internalization during PHD3 loss resulted in receptor hyperactivation under stimulated and growth factor deprived conditions. Importantly, PHD3 physcially associated with the EGFR complex as evidenced by co-immunoprecpitation. Consequently, this extended EGFR activation in PHD3 deficient cells resulted in enhanced downstream activation of EGFR signaling and increased proliferation. Consistent with the interpretation that PHD3 loss is beneficial for tumor growth, we found PHD3 promoter methylation in glioblastoma cell lines, hinting at a epigenetic mechanism to finetune PHD3 expression on top of the hypoxic driven gene regulation. Finally, we demonstrated that PHD3 tumor suppressor function is not restricted to glioblastomas since PHD3 suppression in lung adenocarcinoma accelerated subcutaneous tumor growth. With these findings, we expand the knowledge of PHD3 action from its oxygen sensing role to a regulatory function in growth factor signaling. This clearly discriminates PHD3 from the other isoforms and supports the exclusive tumor suppressor function in glioblastoma. Taken together our results identify a complex role of PHD signaling in cancer and delineate HIF dependent and HIF independent functions of the PHD system. We think that the HIF dependent protective effect of PHD2 and 3 and the HIF independent PHD3 tumor suppressor function are not mutually exclusive, but might be activated according to the heterogeneous intra-tumoral conditions. However, PHD3 hydroxylase activity is dispensable for its HIFindependent tumor suppressor function in glioma. This uncouples PHD3 function from co-factor and co-substrate requirements and allows it to act over a broader physiological range, since its influence on cellular processes is not constrained by the availability of rate limiting factors. It might explain, why the enzymatic independent functions of PHD3 predominate in vivo. Thus, therapeutic modulation of the PHD system to inhibit tumor growth has to be based on these contrasting functions of the PHD system. However, their differential dependence on the hydroxylase activity may facilitate a therapeutic strategy to specifically inhibit or promote the protective versus suppressive functions of the PHD system.
This paper will examine the self-reported division of housework and childcare in Germany and Poland considering the job-related spatial mobility within dual-earner couples who are living in a household together with a partner, using 2007 data from the Job Mobility and Family Lives in Europe Project. We find that men who are spatially mobile for work often report shifting housework to their partners. Polish couples show a stronger tendency toward an egalitarian division of labor than German couples do, especially in terms of childcare. But the central finding of this research is, gender trumps national differences and spatial mobility constraints. Polish and German women, whether mobile for their work or not, report doing the majority of housework and childcare compared to their partners.
The role of gamma oscillatory activity in magnetoencephalogram for auditory memory processing

Recent studies have suggested an important role of cortical gamma oscillatory activity (30-100 Hz) as a correlate of encoding, maintaining and retrieving auditory, visual or tactile information in and from memory. It was shown that these cortical stimulus representations were modulated by attention processes. Gamma-band activity (GBA) occurred as an induced response peaking at approximately 200-300 ms after stimulus presentation. Induced cortical responses appear as non-phase-locked activity and are assumed to reflect active cortical processing rather than passive perception. Induced GBA peaking 200-300 ms after stimulus presentation has been assumed to reflect differences between experimental conditions containing various stimuli. By contrast, the relationship between specific oscillatory signals and the representation of individual stimuli has remained unclear. The present study aimed at the identification of such stimulus-specific gamma-band components. We used magnetoencephalography (MEG) to assess gamma activity during an auditory spatial delayed matching-to-sample task. 28 healthy adults were assigned to one of two groups R and L who were presented with only right- or left-lateralized sounds, respectively. Two sample stimuli S1 with lateralization angles of either 15° or 45° deviation from the midsagittal plane were used in each group. Participants had to memorize the lateralization angle of S1 and compare it to a second lateralized sound S2 presented after an 800-ms delay phase. S2 either had the same or a different lateralization angle as S1. After the presentation of S2, subjects had to indicate whether S1 and S2 matched or not. Statistical probability mapping was applied to the signals at sensor level to identify spectral amplitude differences between 15° and 45° stimuli. We found distinct gamma-band components reflecting each sample stimulus with center frequencies ranging between 59 and 72 Hz in different sensors over parieto-occipital cortex contralateral to the side of stimulation. These oscillations showed maximal spectral amplitudes during the middle 200-300 ms of the delay phase and decreased again towards its end. Additionally, we investigated correlations between the activation strength of the gamma-band components and memory task performance. The magnitude of differentiation between oscillatory components representing 'preferred' and 'nonpreferred' stimuli during the final 100 ms of the delay phase correlated positively with task performance. These findings suggest that the observed gamma-band components reflect the activity of neuronal networks tuned to specific auditory spatial stimulus features. The activation of these networks seems to contribute to the maintenance of task-relevant information in short-term memory.
Fuer die schlechte Prognose von Glioblastompatienten mit einer ueberlebenszeit von 9-15 Monaten (Norden and Wen, 2006) ist vor allem die hohe Invasivitaet dieser Tumore verantwortlich. Nach operativer Entfernung des Haupttumors entstehen aus den verbleibenden invadierten Zellen sekundaere Tumore, die sich mitunter ueber weite Bereiche des Hirns verteilen. Des Weitern sind die hochinvasiven Tumorzellen oft resistent gegen Chemo- und Strahlentherapie (Drappatz et al., 2009; Lefranc et al., 2005). In Maustumormodellen und Pateinten konnte zudem gezeigt werden, dass die neuartige antiangiogenetische Therapie zwar das Tumorwachstum verringert, jedoch die Invasivitaet stark erhoeht. (Norden et al., 2008; Ebos et al., 2009; Paez-Ribes et al., 2009). Ueber die Mechanismen die diese hohen Invasivitaet induzieren, ist bislang nur sehr wenig bekannt. Die durch Reduktion von Blutgefaessen steigende Hypoxie des Tumors foerdert die Expression von Matrix-Metalloproteinasen (MMPs). Dies fuehrt zum Abbau der extrazelluaeren Matrix des umgebenden gesunden Gewebes und beguenstigt dadurch die Tumorzellinvasion (Indelicato et al., 2010; Miyazaki et al., 2008; Shyu et al., 2007). Die Umformung des Aktinzytoskeletts und damit die Mobilitaet von Zellen wird vorwiegend durch ein akkurates Zusammenspeil der Rho GTPasen Rac, Rho und Cdc42, kontrolliert (Ridley et al., 2003). Fuer die Organisation von Axonen im Nervensystem und fuer die Blut- und Lymphgefaessbildung wurde gezeigt, dass die Interaktion der Eph-Rezeptortyrosinkinasen und Ihrer Ephrin-Liganden Signalwege induziert, die in die Regulation dieses Zusammenspiels involviert sind (Egea and Klein, 2007; Makinen et al., 2005; Palmer et al., 2002; Sawamiphak et al., 2010). Des Weiteren zeigt die Analyse der Genloci von Eph-Rezeptoren und Ephrinen in verschieden Hirntumoren eine gehaeufte Deletionen des Ephrin-B2-Gens. Die Quantifizierung von Ephrin-B2 mRNA in diesen Tumoren hat ausserdem ergeben, dass mit zunehmender Malignitaet die Expression von Ephrin-B2 sinkt. Aus diesen Gruenden wurden die Untersuchungen in dieser Arbeit auf die Rolle von Ephrin-B2 anhaengigen Signalwegen in der Glioblastomzellinvasion konzentriert. In einem modifiziertem Boyden-Chamber-Assay konnte gezeigt werden, dass das Ephrin-B2 induzierte EphB4 forward signaling und EphB4 induzierte Ephrin-B2 reverse signaling die Invasivitaet der human Glioblastomzelllinien LN-229, G55 und SNB-19 reduziert. In einem Maustumormodel konnte weiterhin gezeigt werden, dass Ephrin-B2 Knock-Out (KO) Astrozytomzellen, im Vergleich zu Wild-Typ (WT) Zellen, Tumore mit einem groesseren Volumen und einer erhoehten Invasivitaet bilden. Da die Expressionslevel fuer die Ephrin-B2 bindenden Rezeptoren EphA4, EphB1 EphB3 und EphB6 auch im adulten Hirn hoch sind (Hafner et al., 2004), weisen diese in vitro und in vivo Ergebnisse auf eine Tumorsupressorfunktion von Ephrin-B2 hin, die durch repulsive Effekte des Ephrin-B2 reverse signaling vermittelte werden koennten. Dies geht mit Erkenntnissen ueber kolorektale Tumore einher (Batlle et al., 2005). Die in einem Sphaeroid-Invasionsassay mit einer EphB-Rezeptoren freien Umgebung beobachtete verminderte Invasion von Ephrin-B2 WT deutet auf eine zusaetzliche invasionsblockierende Rolle der Ephrin-B2-Eph-Rezeptor Interaktion zwischen benachbarten Tumorzellen hin, wie sie auch in Brusttumoren gefunden wurde (Noren et al., 2006). Es scheint als sei Tumorprogression und Invasion erst moeglich, nachdem die Expression von Ephrin-B2 vermindert wurde. Es konnte weiterhin gezeigt werden, dass in hypoxischen Glioblastomzellen die Ephrin-B2 Expression durch die direkte Bindung des den Transkriptionsfaktors ZEB2 an den Ephrin-B2 Promoter reprimiert wird. In einem Weiteren Maustumormodel konnte gezeigt werden, dass die Blockierung der ZEB2 Expression mittels shRNA und die damit einhergehenden Inhibition der hypoxie induzierten Ephrin-B2 Repression das Wachstum und die Invasivitaet von Glioblastomen verringert. Zusaetzlich wurde gezeigt, dass der Verlust von ZEB2 ausreicht, die durch antiangiogenetische Therapie induzierte stark erhoehte Invasivitaet zu vermeiden. Die in dieser Arbeit gewonnen Erkenntnisse fuehren zu folgendem Modelmechanismus. In kleinen normoxischen Tumoren koennen repulsive Effekte des Ephrin-B2 reverse signalings und EphB forward signalings zwischen Tumorzellen und Zellen des umgebenden Gewebes die Ausbreitung und Invasion des Tumors unterdruecken. Zusaetzlich koennte das Ephrin-B2 induzierte EphB forward signaling zwischen benachbarten Tumorzellen die Mobilitaet der Tumorzellen wie in Brusttumoren inhibieren. Beim Erreichen einer bestimmten Tumorgroesse tritt Hypoxie auf, wodurch HIF-1alpha stabilisiert wird. Dies fuehrt dann zur ZEB2 Expression und leitet die Repression von Ephrin-B2 ein, was wiederum zur erhoehten Tumorzellemobilitaet und im Zusammenspiel mit MMPs zu Invasion fuehren kann. Gleichzeitig werden durch den HIF-induzierten VEGF-Gradienten neue Blutgefaesse rekrutiert. Damit wird der hypoxie-induzierten Invasivitaet entgegengewirkt. Wird mittels antiangiogenetischer Behandlung versucht Tumorprogression entgegenzuwirken, resultiert daraus eine erneut gesteigerte Hypoxie, die dann durch die ZEB2 vermittelte Repression von Ephrin-B2 wieder eine erhoehte Invasivitaet induzieren kann. Das Blockieren der ZEB2 Expression kann dieser durch antiangiogenetischen Behandlung induzierten Invasivitaet entgegenwirken.
Nep1 (Emg1) is a highly conserved nucleolar protein with an essential function in ribosome biogenesis. A mutation in the human Nep1 homolog causes Bowen–Conradi syndrome—a severe developmental disorder. Structures of Nep1 revealed a dimer with a fold similar to the SPOUT-class of RNA-methyltransferases suggesting that Nep1 acts as a methyltransferase in ribosome biogenesis. The target for this putative methyltransferase activity has not been identified yet. We characterized the RNA-binding specificity of Methanocaldococcus jannaschii Nep1 by fluorescence- and NMR-spectroscopy as well as by yeast three-hybrid screening. Nep1 binds with high affinity to short RNA oligonucleotides corresponding to nt 910–921 of M. jannaschii 16S rRNA through a highly conserved basic surface cleft along the dimer interface. Nep1 only methylates RNAs containing a pseudouridine at a position corresponding to a previously identified hypermodified N1-methyl-N3-(3-amino-3-carboxypropyl) pseudouridine (m1acp3-Psi) in eukaryotic 18S rRNAs. Analysis of the methylated nucleoside by MALDI-mass spectrometry, HPLC and NMR shows that the methyl group is transferred to the N1 of the pseudouridine. Thus, Nep1 is the first identified example of an N1-specific pseudouridine methyltransferase. This enzymatic activity is also conserved in human Nep1 suggesting that Nep1 is the methyltransferase in the biosynthesis of m1acp3-Psi in eukaryotic 18S rRNAs.
Rhinoliths are mineralised foreign bodies in the nasal cavity that are a chance finding at anterior rhinoscopy. Undiscovered, they grow appreciably in size and can cause a foul-smelling nasal discharge and breathing problems. Giant nasal stones are now a very rare occurrence, since improved diagnostic techniques, such as endoscopic/microscopic rhinoscopy, now make it possible to identify foreign bodies at an early stage of development. We report the case of a 37-year-old patient who, at the age of 5-6 years, introduced a foreign body, probably a stone, into his right nasal cavity. On presentation, he complained of difficulty in breathing through the right nostril that had persisted for the last 10 years. For the past four years a strong fetid smell from the nose had been apparent to those in his vicinity. Under general anaesthesia, the stone was removed in toto from the right nasal cavity. The possible genesis of the rhinolith is discussed, our case compared with those described in the literature, and possible differential diagnoses are considered.
Until now, the NW Indian Ocean was sparsely covered with coral proxy records, and records from the Maldives Archipelago do not exist. The first such coral proxy record from the central Maldives is presented in this study. It originates from a massive Porites lutea (Quoy and Gaimard, 1833) colony that was sampled March 2007 in the lagoon of Rasdhoo Atoll (4°N/ 73°W), which is located in the central Maldives. The record spans a period of 90 yrs and reaches back to 1917 AD with monthly to bimonthly resolution. This study investigates temporal variations of the skeletal stable oxygen (delta18O) and carbon (delta13C) isotopes, the strontium-to-calcium (Sr/Ca), and the annual extension-rates, and their relationship to historical climate variations 1917-2007. Annual extension-rates show an increase over the 20th century, and are correlated with instrumental sea surface temperatures (SST). The interannual variation of the extension-rates within 2.5-4 years is driven by the El Niño-Southern Oscillation (ENSO). The amount of skeletal extension during the summer months is triggered by variations in the strength of the SW monsoon. Interannual and decadal variability in monsoon current activity (18-19 yrs) and rainfall over India are an expression of the summer monsoon strength. This is the reason why a statistical link between coral extension-rates and precipitation over India can be established. This implies that annual extension-rates in corals can be used as a new proxy for Indian monsoon variability on decadal resolution. The delta18O record exhibits the 20th century warming trend that is influenced by the effect of monsoon-induced cooling. delta18O also reveals interannual ENSO triggered variability, which is due to ENSO-forced variations in SST and sea surface salinity (SSS). A decadal variation at 12-14 yrs cannot be linked to SST variations in the NW Indian Ocean, but with decadal variations of SSS. They could be caused by ENSO- forced variations of the monsoon currents during the mature phase of ENSO teleconnections in the Indian Ocean in boreal winter. The Sr/Ca record does not indicate a significant warming, in spite of the observed SST rise at the sampling site. Changes in seawater Sr/Ca cannot be excluded. Nevertheless, interannual ENSO forcing is still evident. Evidence for the Pacific Decadal Oscillation (PDO) is found during 1917-1955. Afterwards, the Sr/Ca data indicate the disappearance of PDO forcing. By the combination of Sr/Ca and delta18O it is possible to detect ~80% of historical El Niño and La Niña events at the sample site. This study confirms the notion that interannual to multi-decadal climate fluctuations in the Pacific play a crucial role for climate variability in the Indian Ocean.
The QCD phase diagram as a function of temperature, T, and chemical potential for baryon
number, mB, is still unknown today, due to the sign problem, which prohibits direct Monte Carlo
simulations for non-vanishing baryon density. Investigations in models sharing chiral symmetry
with QCD predict a phase diagram, in which the transition corresponds to a smooth crossover at
zero density, but which is strengthened by chemical potential to turn into a first order transition
beyond some second order critical point. This contribution reviews the lattice evidence in favour
and against the existence of a critical point.

The physical and functional borders of transit peptide-like sequences in secondary endosymbionts

Background: Plastids rely on protein supply by their host cells. In plastids surrounded by two membranes (primary plastids) targeting of these proteins is facilitated by an N-terminal targeting signal, the transit peptide. In secondary plastids (surrounded by three or four membranes), transit peptide-like regions are an essential part of a bipartite topogenic signal sequence (BTS), and generally found adjacent to a N-terminally located signal peptide of the plastid pre-proteins. As in primary plastids, for which no wealth of functional information about transit peptide features exists, the transit peptide-like regions used for import into secondary ones show some common features only, which are also poorly characterised. Results: We modified the BTS (in the transit peptide-like region) of the plastid precursor fucoxanthin-chlorophyll a/c binding protein D (FcpD) fused to GFP as model substrate for the characterisation of pre-protein import into the secondary plastids of diatoms. Thereby we show that (i) pre-protein import is highly charge dependent. Positive net charge is necessary for transport across the plastid envelope, but not across the periplastid membrane. Acidic net charge perturbs pre-protein import within the ER. Moreover, we show that (ii) the mature domain of the pre-protein can provide intrinsic transit peptide functions. Conclusions: Our results indicate important characteristics of targeting signals of proteins imported into secondary plastids surrounded by four membranes. In addition, we show a self-targeting mechanism, in which the mature protein domain contributes to the transit peptide function. Thus, this phenomenon lowers the demand for pre-sequences evolved during the course of endosymbiosis.
Background: New drugs are constantly sought after to improve the survival of patients with malignant gliomas. The ideal substance would selectively target tumor cells without eliciting toxic side effects. Here, we report on the anti-proliferative, anti-migratory, and anti-invasive properties of the natural, nontoxic compound Curcumin observed in five human glioblastoma (GBM) cell lines in vitro. Methods: We used monolayer wound healing assays, modified Boyden chamber trans-well assays, and cell growth assays to quantify cell migration, invasion, and proliferation in the absence or presence of Curcumin at various concentrations. Levels of the transcription factor phospho-STAT3, a potential target of Curcumin, were determined by sandwich-ELISA. Subsequent effects on transcription of genes regulating the cell cycle were analyzed by quantitative real-time PCR. Effects on apoptosis were determined by caspase assays. Results: Curcumin potently inhibited GBM cell proliferation as well as migration and invasion in all cell lines contingent on dose. Simultaneously, levels of the biologically active phospho-STAT3 were decreased and correlated with reduced transcription of the cell cycle regulating gene c-Myc and proliferation marking Ki-67, pointing to a potential mechanism by which Curcumin slows tumor growth. Conclusions: Curcumin is part of the diet of millions of people every day and is without known toxic side effects. Our data show that Curcumin bears anti-proliferative, anti-migratory, and anti-invasive properties against GBM cells in vitro. These results warrant further in vivo analyses and indicate a potential role of Curcumin in the treatment of malignant gliomas.
The KADoNiS (Karlsruhe Astrophysical Database of Nucleosynthesis in Stars) project is an online
database (www.kadonis.org) for cross sections relevant to the s-process and the p-process.
The first version was an updated sequel to the previous Bao et al. [1] compilations from 1987
and 2000 for (n; g) cross sections relevant to Big Bang and s-process nucleosynthesis. The first
update, KADoNiS v0.2, was published in 2006 [2]. It contained mainly Maxwellian averaged
(n; g) cross sections relevant to the s-process, and some experimental charged particle induced
reaction relevant to the p-process. After that a second update was presented in 2009 [3].
Recently, we started to collect and review all existing experimental data relevant for p-process
nucleosynthesis and to provide a user-friendly database based on the KADoNiS framework. The
p-process part of the KADoNiS database is currently being extended and will include all available
experimental data from (p; g), (p;n), (p;a), (a,g), (a;n) and (a; p) reactions in or close to the
respective Gamow window.
The pathophysiology of schizophrenia is still poorly understood. Investigating the neurophysiological correlates of cognitive dysfunction with functional neuroimaging techniques such as electroencephalography (EEG) and functional magnetic resonance imaging (fMRI) is widely considered to be a possible solution for this problem. Working memory impairment is one of the most prominent cognitive impairments found in schizophrenia. Working memory can be divided into a number of component processes, encoding, maintenance and retrieval. They appear to be differentially affected in schizophrenia, but little is known about the neurophysiological disturbances which contribute to deficits in these component processes. The aim of this dissertation was to elucidate the neurophysiological underpinnings of the component processes of working memory and their disturbance in schizophrenia. In the first study the the neurophysiological substrates of visual working memory capacity limitations were investigated during encoding, maintenance and retrieval in 12 healthy subjects using event-related fMRI. Subjects had to encode up to four abstract visual shapes and maintain them in working memory for 12 seconds. Afterwards a test stimulus was presented, which matched one of the previously shown shapes in fifty percent of the trials. A bilateral inverted U-shape pattern of BOLD activity with increasing memory load in areas closely linked with selective attention, i.e. the frontal eye fields and areas around the intraparietal sulcus, was observed already during encoding. The increase of the number of stored items from memory load three to memory load four in these regions was negatively correlated with the increase of BOLD activity from memory load three to memory load four. These results point to a crucial role of attentional processes for the limited capacity of working memory. In the second study, the contribution of early perceptual processing deficits during encoding and retrieval to working memory dysfunction was investigated in 17 patients with schizophrenia and 17 healthy control subjects using EEG and event-related fMRI. A slightly modified version of the working memory task used in the fist study was employed. Participants only had to encode and maintain up to three items. In patients the amplitude of the P1 event-related potential was significantly reduced already during encoding in all memory load conditions. Similarly, BOLD activity in early visual areas known to generate the P1 was significantly reduced in patients. In controls, a stronger P1 amplitude increase with increasing memory load predicted better performance. These findings indicate that in addition to later memory related processing stages early visual processing is disturbed in schizophrenia and contributes to working memory dysfunction by impairing the encoding of information. In the third study, which was based on the same data set as the second study, cortical activity and functional connectivity in 17 patients with schizophrenia and 17 to healthy control subjects during the working memory encoding, maintenance and retrieval was investigated using event-related fMRI. Patients had reduced working memory capacity. During encoding activation in the left ventrolateral prefrontal cortex and extrastriate visual cortex was reduced in patients but positively correlated with working memory capacity in controls. During early maintenance patients switched from hyper- to hypoactivation with increasing memory load in a fronto-parietal network which included left dorsolateral prefrontal cortex. During retrieval right ventrolateral prefrontal hyperactivation was correlated with encoding-related hypoactivation of left ventrolateral prefrontal cortex in patients. Cortical dysfunction in patients during encoding and retrieval was accompanied by abnormal functional connectivity between fronto-parietal and visual areas. These findings indicate a primary encoding deficit in patients caused by a dysfunction of prefrontal and visual areas. The findings of these studies suggest that isolating the component processes of working memory leads to more specific markers of cortical dysfunction in schizophrenia, which had been obscured in previous studies. This approach may help to identify more reliable biomarkers and endophenotypes of schizophrenia.
Introduction: Acute lung injury (ALI) is an inflammatory disorder of pulmonary or extrapulmonary origin. We have previously demonstrated that netrin-1 dampens murine ALI, and in an attempt to advance this finding into future clinical practice we evaluated whether netrin-1 would reduce alveolar inflammation during porcine ALI. Methods: This was a controlled in vivo experimental study in pigs. We induced ALI through lipoploysaccharide (LPS) infusion (50 micro g/kg) for 2 hours. Following this, we exposed animals to either vehicle, intravenous netrin-1 (netrin-1 i.v.) or inhaled netrin-1 (netrin-1 inh.). Serum samples and bronchoalveolar lavage (BAL) were obtained to determine levels of tumor necrosis factor-alpha (TNF-alpha), interleukin (IL)-1beta, interleukin-6 and interleukin-8 at baseline and 6 hours following treatment. Myeloperoxidase activity (MPO) and protein levels were determined in the BAL, and tissue samples were obtained for histological evaluation. Finally, animals were scanned with spiral CT. Results: Following LPS infusion, animals developed acute pulmonary injury. Serum levels of TNF-alpha and IL-6 were significantly reduced in the netrin-1 i.v. group. BAL demonstrated significantly reduced cytokine levels 6 hours post-netrin-1 treatment (TNF-alpha: vehicle 633 ± 172 pg/ml, netrin-1 i.v. 84 ± 5 pg/ml, netrin-1 inh. 168 ± 74 pg/ml; both P < 0.05). MPO activity and protein content were significantly reduced in BAL samples from netrin-1-treated animals. Histological sections confirmed reduced inflammatory changes in the netrin-1-treated animals. Computed tomography corroborated reduced pulmonary damage in both netrin-1-treated groups. Conclusions: We conclude that treatment with the endogenous anti-inflammatory protein netrin-1 reduces pulmonary inflammation during the initial stages of ALI and should be pursued as a future therapeutic option.
Theory building is not only underdeveloped in IT services management research, but in
general in IS. Given the paradigm shift that comes from the development away from a
networked economy towards a network economy, the lack of spending enough attention to
theorizing in IS becomes even more obvious. In the light of other "megatrends" in IS
research, such as the increasing professionalization and use of statistical methods and the
exploitation of extremely large sets of data (often harvested from social media sites), we
might lose interest in theorizing in the presence of the tremendous amount of available
empirical data. In this position paper, the author advocates that services science researchers
should focus on rigor and relevance in their research approaches.
Die Maillard-Reaktion findet während der Lagerung und thermischen Verarbeitung von Lebensmitteln zwischen den darin enthaltenen Proteinen und reduzierenden Kohlehydraten statt. Als Ergebnis der Reaktion entstehen sogenannte advanced glycation end products (AGEs), Protein-Derivate mit Glykierungs-Strukturen. Da Lebensmittel vor dem Verzehr häufig erhitzt werden, ist der Einfluss von AGEs auf die Pathogenese von Nahrungsmittelallergien von großem Interesse. Die Maillard-Reaktion könnte zur Bildung von neuen, für die Pathogenese der Nahrungsmittelallergie relevanten, Immunepitopen beitragen. Das Ziel dieser Arbeit war es, den Einfluss der Maillard-Reaktion auf die T-Zell-Immunogenität, die Antigenität und die von beiden Eigenschaften abhängige Allergenität von Nahrungsmittelallergenen zu untersuchen. Zunächst wurde der Einfluss der Maillard-Reaktion auf die T-Zell-Immunogenität von Ovalbumin (OVA), einem Allergen des Hühnereiweißes, untersucht. Dafür wurde glykiertes OVA (AGE-OVA) hergestellt indem das Protein zusammen mit Glukose erhitzt wurde. In dieser Arbeit konnte zum ersten Mal gezeigt werden, dass ein AGE-Derivat eines Lebensmittelallergens eine höhere T-Zellen-Immunogenität besitzt, als sein natives Gegenstück. Die Aktivierung und Proliferation von CD4+ T-Zellen durch AGE-OVA wurde in vitro durch Co-Kultivierung der T-Zellen mit dendritischen Zellen (DZ) untersucht. DZ sind professionelle Antigen- präsentierende Zellen, welche im Pathomechanismus der Allergie eine wichtige Rolle spielen. Im Vergleich zu nativen OVA und OVA welches ohne Glukose erhitzt wurde, führte die Stimulierung mit AGE-OVA zu einer deutlich erhöhten Aktivierung von OVA-spezifischen CD4+ T-Zellen. Damit DZ T-Zellen aktivieren können, muss das Allergen zunächst durch die DZ aufgenommen werden. In der vorliegenden Arbeit wurde gezeigt, dass die Aufnahme von AGE-OVA wesentlich höher war als die der Kontrollen. Außerdem konnte der scavenger receptor class A type I and II (SR-AI/II) als einer der hauptverantwortlichen Rezeptoren für die Aufnahme von AGE-OVA identifiziert werden. Zusammenfassend lässt sich aus den Ergebnissen dieser Arbeit die Hypothese aufstellen, dass die Glykierung von OVA eine erhöhte Assoziation des Allergens mit SR-AI/II ermöglicht, welche zu einer verstärkten Aufnahme des Allergens durch die DZ führt. Dadurch können mehr Peptide des Allergens an MHC II gebunden und auf der Zelloberfläche präsentiert werden. Das wiederum führt zur beobachteten stärkeren OVA-spezifischen CD4+ T-Zell-Aktivierung durch AGE-OVA. Als nächstes wurde die T-Zell-Immunogenität und Antigenität von AGE-OVA in vivo in einem Mausmodel untersucht. Es zeigte sich, dass AGE-OVA auch in vivo im Vergleich zu den nicht glykierten OVA-Formen eine erhöhte T-Zell-Immunogenität besitzt. Des weiteren führte die Immunisierung mit AGE-OVA zu einer erhöhten Produktion von IgE-Antikörpern. Somit wurde in dieser Arbeit gezeigt, dass AGE-OVA in vivo nicht nur eine erhöhte CD4+ T-Zell-Immunogenität besitzt, sondern auch eine höhere Antigenität hat als natives und ohne Glukose erhitztes OVA. Diese Ergebnisse harmonieren gut miteinander da CD4+ T-Zellen eine zentrale Rolle in der Aktivierung von B-Zellen und der IgE-Produktion durch selbige Zellen spielen. IgE-Antikörper besitzen eine essentielle Funktion beim Auslösen der klinischen Symptomatik der Allergie. Zusammenfassend lässt deshalb sagen, dass die Maillard-Reaktion die Allergenität von OVA erhöhen könnte. Zum Schluss wurden noch die immunstimulatorischen Eigenschaften des Erdnussallergens (AGE)-Ara h 2 untersucht. Da Erdnüsse häufig ernsthafte allergische Reaktionen hervorrufen und selten roh verzehrt werden, war es vom großen Interesse den Einfluss der Maillard-Reaktion auf Immunogenität und Antigenität von rekombinanten Ara h 2 (rAra h 2) zu untersuchen. Es zeigte sich, dass die Glykierung von rAra h 2 durch die Maillard-Reaktion die T-Zellen-Immunogenität, als auch die Antigenität des Allergens reduziert. Abschließend lässt sich sagen, dass die Maillard-Reaktion die allergenen Eigenschaften von Lebensmittelallergenen erheblich beeinflusst indem es die T-Zell-Immunogenität des Allergens verändert. Die Mechanismen welche die T-Zell-Immunogenität beeinflussen wurden hier näher untersucht. Wenn die Glykierung nicht die Bindung der T-Zellen- und/oder B-Zellen-Rezeptoren inhibiert, wird die Allergen-spezifische CD4+ T-Zell-Aktivierung und die davon abhängige IgE-Produktion dadurch erhöht, dass das glykierte Allergen durch DZ verstärkt über SR-AI/II aufgenommen wird. Die vorliegende Arbeit liefert wertvolle Information über die Allergenität von Proteinen die durch die Maillard-Reaktion modifiziert wurden and trägt dazu bei die Mechanismen von Nahrungsmittelallergien besser zu verstehen.
From the very outset of European expansion, scholars have been preoccupied with the impact of proselytization and colonization on non-European societies. Anthropologists such as Margaret Mead and Bronislaw Malinowski, who witnessed these processes at the beginning of the twentieth century while at the same time benefitting from the colonial structure, were convinced that the autochthonous societies could not possibly withstand the onslaught of the dominant European cultures, and thus were doomed to vanish in the near future. The fear of losing their object of research, which had just recently been discovered, hung above the heads of the scholars like a sword of Damocles ever since the establishment of anthropology as a discipline. They felt hurried to document what seemed to be crumbling away. Behind these fears there was the notion that the indigenous cultures were comparatively static entities that had existed untouched by any external influences for many centuries, or even millennia, and were unable to change. This idea was shared by proponents of other disciplines; in religious studies, for example, up to the late 1980s the view prevailed that the contact between the great world religions and the belief systems of small, autochthonous societies doomed the latter to extinction. However, more recent studies have shown that this assumption, according to which indigenous peoples have not undergone any changes in the course of history, is untenable. It became apparent that groups supposedly living in isolation have extensive contact networks, and that migration, trade, and conquest are not privileges of modern times. Myths and oral traditions bore witness of journeys to faraway regions, new settlements founded in unknown territories, or the arrival of victorious foreigners who introduced new ways and customs and laid claim to a place of their own within society.
The impact of shift work induced chronic circadian disruption on IL-6 and TNF-α immune responses

Aim: Sleep disturbances induce proinflammatory immune responses, which might increase cardiovascular disease risk. So far the effects of acute sleep deprivation and chronic sleep illnesses on the immune system have been investigated. The particular impact of shift work induced chronic circadian disruption on specific immune responses has not been addressed so far.
Methods: Pittsburgh-Sleep-Quality-Index (PSQI) questionnaire and blood sampling was performed by 225 shift workers and 137 daytime workers. As possible markers the proinflammatory cytokines IL-6 and TNF-alpha and lymphocyte cell count were investigated. A medical examination was performed and biometrical data including age, gender, height, weight, waist and hip circumference and smoking habits were collected by a structured interview.
Results: Shift workers had a significantly higher mean PSQI score than day workers (6.73 vs. 4.66; p < 0.001). Day workers and shift workers had similar serum levels of IL-6 (2.30 vs. 2.67 resp.; p = 0.276), TNF-alpha (5.58 vs. 5.68, resp.; p = 0.841) or lymphocytes count (33.68 vs. 32.99, resp.; p = 0.404). Furthermore there were no differences in cytokine levels (IL-6 p = 0.761; TNF-alpha p = 0.759) or lymphocyte count (p = 0.593) comparing the sleep quality within the cohorts. When this calculation of sleep quality was stratified by shift and day workers irrespective of their sleep quality day workers and shift workers had similar serum levels of IL-6, TNF-alpha or lymphocytes count. Multiple linear regression analysis showed a significant correlation of lymphocytes count and smoking habits.
Conclusion: Shift work induces chronic sleep debt. Our data reveals that chronic sleep debt might not always lead to an activation of the immune system, as we did not observe differences in lymphocyte count or level of IL-6 or TNF-alpha serum concentration between shift workers and day workers. Therefore chronic sleep restriction might be eased by a long-term compensating immune regulation which (in healthy) protects against an overstimulation of proinflammatory immune mechanisms and moderates metabolic changes, as they are known from short-term sleep deprivation or sleep related breathing disorders.
Disruptive behaviour disorders are reflected by a great variety of symptoms ranging from impulsive-hot tempered quarrels to purposeful and goal directed acts of cruelty. A growing body of data indicates that there are neurobiological factors that increase the risk for developing disruptive behaviour disorders. In this review, we give a broad overview of recent studies investigating physiological, neural, genetic factors, and specific neurotransmitter systems. We also discuss the impact of psychosocial risk and consider the effects of gene-environment interactions. Due to the heterogeneity of disruptive behaviour disorders, it is concluded that specific subtypes of disruptive behaviour should be considered both in terms their biological basis and in regard to specific treatment needs.
Emissions of sulphur hexafluoride (SF6), one of the strongest greenhouse gases on a per molecule basis, are targeted to be collectively reduced under the Kyoto Protocol. Because of its long atmospheric lifetime (estimated as 800 to 3200 years), the accumulation of SF6 in the atmosphere is a direct measure of its global emissions. Examination of our extended data set of globally distributed high-precision SF6 observations shows an increase in SF6 abundance from near zero in the 1970s to a global mean of 6.7 ppt by the end of 2008. In-depth evaluation of our long-term data records shows that the global source of SF6 decreased after 1995, most likely due to SF6 emission reductions in industrialised countries, but increased again after 1998. By subtracting those emissions reported by Annex I countries to the United Nations Framework Convention of Climatic Change (UNFCCC) from our observation-inferred SF6 source leaves a surprisingly large gap of more than 70–80% of non-reported SF6 emissions in the last decade. This suggests a strong under-estimation of emissions in Annex I countries and underlines the urgent need for independent atmospheric verification of greenhouse gases emissions accounting.
