Background: Zolpidem is a non-benzodiazepine hypnotic agent which has been shown to be effective in inducing and maintaining sleep in adults and is one of the most frequently prescribed hypnotics in the world. For drugs that are used to treat sleeping disorders, the time to reach the maximum concentration (Tmax) of the drug in plasma is important to achieving a fast onset of action and this must be maintained when switching from one product to another.
Objectives: The main objective of the present work was to create a PBPK/PD model for zolpidem and establish a clinically relevant “safe space” for dissolution of zolpidem from the commercial immediate release (IR) formulation. A second objective was to analyze literature pharmacokinetic data to verify the negative food effect ascribed to zolpidem and consider its ramifications in terms of the “safe space” for dissolution.
Methods: Using dissolution, pharmacokinetic and pharmacodynamic data, an integrated PBPK/PD model for immediate release zolpidem tablets was constructed in Simcyp®. This model was used to identify the clinically relevant dissolution specifications necessary to ensure efficacy.
Results: According to the simulations, as long as 85% of the drug is released in 45 minutes or less, the impact on the PK and PD profiles of zolpidem would be minimal. According to the FDA, the drug has to dissolve from the test and reference products at a similar rate and to an extent of 85% in not more than 30 minutes to pass bioequivalence via the BCS-biowaiver test. Thus, the BCS-biowaiver specifications are somewhat more stringent than the “safe space” based on the PBPK/PD model. Published data from fasted and fed state pharmacokinetic studies suggest but do not prove a negative food effect of zolpidem.
Conclusions: A PBPK/PD model indicates that current BCS biowaiver criteria are more restrictive for immediate release zolpidem tablets than they need to be. In view of the close relationship between PK and PD, it remains advisable to avoid taking zolpidem tablets with or immediately after a meal, as indicated by the Stilnox® labeling.
O presente objeto de pesquisa busca proceder ao estudo e identificação dos traços essenciais envolvidos na abordagem teórica das relações sociais e políticas trazidas na obra O Direito da Liberdade do filósofo alemão Axel Honneth. Faz-se uma análise da influência hegeliana sobre o conceito de liberdade, assim como dos fatores relacionados com o suprimento das carências subjetivas, mediadas pelas diferentes “esferas” sociais. Honneth, assim, procura trazer à tona a compreensão de um novo modelo de liberdade advindo da Filosofia do Direito de Hegel, o qual se distingue substancialmente dos modelos tradicionais. O autor busca evidenciar a limitação das teorias da justiça de tradição liberal, invocando a necessidade de uma visão integrada das relações sociais experimentadas nas esferas referidas por Hegel, concebendo-se uma experiência concreta de liberdade social. Nesse sentido, evidencia-se o caráter interdisciplinar e emancipatório do método de reconstrução normativa como base teórica para a justificação pública nas sociedades modernas.
Background: While systemic inflammation is recognized as playing a central role in the pathogenesis of organ failures in patients with liver cirrhosis, less is known about its relevance in the development of classical hepatic decompensation. Aim: To characterize the relationship between systemic inflammation, hemodynamics, and anemia with decompensation of liver cirrhosis. Methods: This is a post-hoc analysis of a cohort study of outpatients with advanced liver fibrosis or cirrhosis. Results: Analysis included 338 patients of whom 51 patients (15%) were hospitalized due to decompensation of liver cirrhosis during a median follow-up time of six months. In univariate analysis, active alcoholism (p = 0.002), model of end-stage liver disease (MELD) score (p = 0.00002), serum IL-6 concentration (p = 0.006), heart rate (p = 0.03), low arterial blood pressure (p < 0.05), maximal portal venous flow (p = 0.008), and low hemoglobin concentration (p < 0.00001) were associated with hospitalization during follow-up. Multivariate analysis revealed an independent association of low hemoglobin (OR = 0.62, 95% CI = 0.51–0.78, p = 0.001) and serum IL-6 concentration (OR = 1.02, 95% CI = 1.01–1.04, p = 0.03)—but not of hemodynamic parameters—with hepatic decompensation. An inverse correlation between hemoglobin concentration and portal venous flow (R = −0.362, p < 0.0001) was detected for the non-hospitalized patients. Accuracy of baseline hemoglobin levels for predicting hospitalization (AUC = 0.84, p < 0.000001) was high. Conclusion: Anemia and systemic inflammation, rather than arterial circulatory dysfunction, are strong and independent predictors of hepatic decompensation in outpatients with liver cirrhosis.
Background: Sepsis frequently occurs after major trauma and is closely associated with dysregulations in the inflammatory/complement and coagulation system. Thrombin-activatable fibrinolysis inhibitor (TAFI) plays a dual role as an anti-fibrinolytic and anti-inflammatory factor by downregulating complement anaphylatoxin C5a. The purpose of this study was to investigate the association between TAFI and C5a levels and the development of post-traumatic sepsis. Furthermore, the predictive potential of both TAFI and C5a to indicate sepsis occurrence in polytraumatized patients was assessed. Methods: Upon admission to the emergency department (ED) and daily for the subsequent ten days, circulating levels of TAFI and C5a were determined in 48 severely injured trauma patients (injury severity score (ISS) ≥ 16). Frequency matching according to the ISS in septic vs. non-septic patients was performed. Trauma and physiologic characteristics, as well as outcomes, were assessed. Statistical correlation analyses and cut-off values for predicting sepsis were calculated. Results: Fourteen patients developed sepsis, while 34 patients did not show any signs of sepsis (no sepsis). Overall injury severity, as well as demographic parameters, were comparable between both groups (ISS: 25.78 ± 2.36 no sepsis vs. 23.46 ± 2.79 sepsis). Septic patients had significantly increased C5a levels (21.62 ± 3.14 vs. 13.40 ± 1.29 ng/mL; p < 0.05) and reduced TAFI levels upon admission to the ED (40,951 ± 5637 vs. 61,865 ± 4370 ng/mL; p < 0.05) compared to the no sepsis group. Negative correlations between TAFI and C5a (p = 0.0104) and TAFI and lactate (p = 0.0423) and positive correlations between C5a and lactate (p = 0.0173), as well as C5a and the respiratory rate (p = 0.0266), were found. In addition, correlation analyses of both TAFI and C5a with the sequential (sepsis-related) organ failure assessment (SOFA) score have confirmed their potential as early sepsis biomarkers. Cut-off values for predicting sepsis were 54,857 ng/mL for TAFI with an area under the curve (AUC) of 0.7550 (p = 0.032) and 17 ng/mL for C5a with an AUC of 0.7286 (p = 0.034). Conclusion: The development of sepsis is associated with early decreased TAFI and increased C5a levels after major trauma. Both elevated C5a and decreased TAFI may serve as promising predictive factors for the development of sepsis after polytrauma.
Low-to-moderate quality meta-analytic evidence shows that motor control stabilisation exercise (MCE) is an effective treatment of non-specific low back pain. A possible approach to overcome the weaknesses of traditional meta-analyses would be that of a prospective meta-analyses. The aim of the present analysis was to generate high-quality evidence to support the view that motor control stabilisation exercises (MCE) lead to a reduction in pain intensity and disability in non-specific low back pain patients when compared to a control group. In this prospective meta-analysis and sensitivity multilevel meta-regression within the MiSpEx-Network, 18 randomized controlled study arms were included. Participants with non-specific low back pain were allocated to an intervention (individualized MCE, 12 weeks) or a control group (no additive exercise intervention). From each study site/arm, outcomes at baseline, 3 weeks, 12 weeks, and 6 months were pooled. The outcomes were current pain (NRS or VAS, 11 points scale), characteristic pain intensity, and subjective disability. A random effects meta-analysis model for continuous outcomes to display standardized mean differences between intervention and control was performed, followed by sensitivity multilevel meta-regressions. Overall, 2391 patients were randomized; 1976 (3 weeks, short-term), 1740 (12 weeks, intermediate), and 1560 (6 months, sustainability) participants were included in the meta-analyses. In the short-term, intermediate and sustainability, moderate-to-high quality evidence indicated that MCE has a larger effect on current pain (SMD = −0.15, −0.15, −0.19), pain intensity (SMD = −0.19, −0.26, −0.26) and disability (SMD = −0.15, −0.27, −0.25) compared with no exercise intervention. Low-quality evidence suggested that those patients with comparably intermediate current pain and older patients may profit the most from MCE. Motor control stabilisation exercise is an effective treatment for non-specific low back pain. Sub-clinical intermediate pain and middle-aged patients may profit the most from this intervention.
In the course of a growing start-up market and strongly increasing investment volume, investors try to predict the success of a business as precisely as possible in advance. However, when assessing the personality of the founder or founding team, they still rely far too often on their gut feeling, thereby reducing the quality of their decisions. Our study therefore aimed at investigating whether there are any relationships between the founders' personality traits and their performance and thus justifying the need for more targeted and optimized diagnostics in the field of founder personality. With a total of 141 founders, clear correlations between personality traits (conscientiousness, emotional stability) and performance could be demonstrated in the present study. In addition, it became evident that perceived stress is also related to the founders’ personality (emotional stability negative, conscientiousness positive) and in turn has a negative effect on performance. Our findings contribute to raising awareness of the importance of personality as a predictor of founders' performance, improving decision-making, and, in the long run, replacing gut feeling as an inappropriate assessment criterion of investors.
Background: More than 170 species of tabanids are known in Europe, with many occurring only in limited areas or having become very rare in the last decades. They continue to spread various diseases in animals and are responsible for livestock losses in developing countries. The current monitoring and recording of horseflies is mainly conducted throughout central Europe, with varying degrees of frequency depending on the country. To the detriment of tabanid research, little cooperation exists between western European and Eurasian countries.
Methods: For these reasons, we have compiled available sources in order to generate as complete a dataset as possible of six horsefly species common in Europe. We chose Haematopota pluvialis, Chrysops relictus, C. caecutiens, Tabanus bromius, T. bovinus and T. sudeticus as ubiquitous and abundant species within Europe. The aim of this study is to estimate the distribution, land cover usage and niches of these species. We used a surface-range envelope (SRE) model in accordance with our hypothesis of an underestimated distribution based on Eurocentric monitoring regimes.
Results: Our results show that all six species have a wide range in Eurasia, have a broad climatic niche and can therefore be considered as widespread generalists. Areas with modelled habitat suitability cover the observed distribution and go far beyond these. This supports our assumption that the current state of tabanid monitoring and the recorded distribution significantly underestimates the actual distribution. Our results show that the species can withstand extreme weather and climatic conditions and can be found in areas with only a few frost-free months per year. Additionally, our results reveal that species prefer certain land-cover environments and avoid other land-cover types.
Conclusions: The SRE model is an effective tool to calculate the distribution of species that are well monitored in some areas but poorly in others. Our results support the hypothesis that the available distribution data underestimate the actual distribution of the surveyed species.
Reduced external knee adduction moments in the second half of stance after total hip replacement have been reported in hip osteoarthritis patients. This reduction is thought to shift the load from the medial to the lateral knee compartment and as such increase the risk for knee osteoarthritis. The knee adduction moment is a surrogate for the load distribution between the medial and lateral compartments of the knee and not a valid measure for the tibiofemoral contact forces which are the result of externally applied forces and muscle forces. The purpose of this study was to investigate whether the distribution of the tibiofemoral contact forces over the knee compartments in unilateral hip osteoarthritis patients 1 year after receiving a primary total hip replacement differs from healthy controls. Musculoskeletal modeling on gait was performed in OpenSim using the detailed knee model of Lerner et al. (2015) for 19 patients as well as for 15 healthy controls of similar age. Knee adduction moments were calculated by the inverse dynamics analysis, medial and lateral tibiofemoral contact forces with the joint reaction force analysis. Moments and contact forces of patients and controls were compared using Statistical Parametric Mapping two-sample t-tests. Knee adduction moments and medial tibiofemoral contact forces of both the ipsi- and contralateral leg were not significantly different compared to healthy controls. The contralateral leg showed 14% higher medial tibiofemoral contact forces compared to the ipsilateral (operated) leg during the second half of stance. During the first half of stance, the lateral tibiofemoral contact force of the contralateral leg was 39% lower and the ratio 32% lower compared to healthy controls. In contrast, during the second half of stance the forces were significantly higher (39 and 26%, respectively) compared to healthy controls. The higher ratio indicates a changed distribution whereas the increased lateral tibiofemoral contact forces indicate a higher lateral knee joint loading in the contralateral leg in OA patients after total hip replacement (THR). Musculoskeletal modeling using a detailed knee model can be useful to detect differences in the load distribution between the medial and lateral knee compartment which cannot be verified with the knee adduction moment.
Objective: Many patients with localized prostate cancer (PCa) do not immediately undergo radical prostatectomy (RP) after biopsy confirmation. The aim of this study was to investigate the influence of “time-from-biopsy-to- prostatectomy” on adverse pathological outcomes.
Materials and Methods: Between January 2014 and December 2019, 437 patients with intermediate- and high risk PCa who underwent RP were retrospectively identified within our prospective institutional database. For the aim of our study, we focused on patients with intermediate- (n = 285) and high-risk (n = 151) PCa using D'Amico risk stratification. Endpoints were adverse pathological outcomes and proportion of nerve-sparing procedures after RP stratified by “time-from-biopsy-to-prostatectomy”: ≤3 months vs. >3 and < 6 months. Medians and interquartile ranges (IQR) were reported for continuously coded variables. The chi-square test examined the statistical significance of the differences in proportions while the Kruskal-Wallis test was used to examine differences in medians. Multivariable (ordered) logistic regressions, analyzing the impact of time between diagnosis and prostatectomy, were separately run for all relevant outcome variables (ISUP specimen, margin status, pathological stage, pathological nodal status, LVI, perineural invasion, nerve-sparing).
Results: We observed no difference between patients undergoing RP ≤3 months vs. >3 and <6 months after diagnosis for the following oncological endpoints: pT-stage, ISUP grading, probability of a positive surgical margin, probability of lymph node invasion (LNI), lymphovascular invasion (LVI), and perineural invasion (pn) in patients with intermediate- and high-risk PCa. Likewise, the rates of nerve sparing procedures were 84.3 vs. 87.4% (p = 0.778) and 61.0% vs. 78.8% (p = 0.211), for intermediate- and high-risk PCa patients undergoing surgery after ≤3 months vs. >3 and <6 months, respectively. In multivariable adjusted analyses, a time to surgery >3 months did not significantly worsen any of the outcome variables in patients with intermediate- or high-risk PCa (all p > 0.05).
Conclusion: A “time-from-biopsy-to-prostatectomy” of >3 and <6 months is neither associated with adverse pathological outcomes nor poorer chances of nerve sparing RP in intermediate- and high-risk PCa patients.
MicroRNAs (miRNAs) have emerged as critical posttranscriptional regulators of the immune system, including function and development of regulatory T (Treg) cells. Although this critical role has been firmly demonstrated through genetic models, key mechanisms of miRNA function in vivo remain elusive. Here, we review the role of miRNAs in Treg cell development and function. In particular, we focus on the question what the study of miRNAs in this context reveals about miRNA biology in general, including context-dependent function and the role of individual targets vs. complex co-targeting networks. In addition, we highlight potential technical pitfalls and state-of-the-art approaches to improve the mechanistic understanding of miRNA biology in a physiological context.
Despite the success of immune checkpoint blockade in cancer, the number of patients that benefit from this revolutionary treatment option remains low. Therefore, efforts are being undertaken to sensitize tumors for immune checkpoint blockade, which includes combining immune checkpoint blocking agents such as anti-PD-1 antibodies with standard of care treatments. Here we report that a combination of chemotherapy (doxorubicin) and immune checkpoint blockade (anti-PD-1 antibodies) induces superior tumor control compared to chemotherapy and immune checkpoint blockade alone in the murine autochthonous polyoma middle T oncogene-driven (PyMT) mammary tumor model. Using whole transcriptome analysis, we identified a set of genes that were upregulated specifically upon chemoimmunotherapy. This gene signature and, more specifically, a condensed four-gene signature predicted favorable survival of human mammary carcinoma patients in the METABRIC cohort. Moreover, PyMT tumors treated with chemoimmunotherapy contained higher levels of cytotoxic lymphocytes, particularly natural killer cells (NK cells). Gene set enrichment analysis and bead-based ELISA measurements revealed increased IL-27 production and signaling in PyMT tumors upon chemoimmunotherapy. Moreover, IL-27 signaling improved NK cell cytotoxicity against PyMT cells in vitro. Taken together, our data support recent clinical observations indicating a benefit of chemoimmunotherapy compared to monotherapy in breast cancer and suggest potential underlying mechanisms.
Although cyclophosphamide (CP) has been used successfully in the clinic for over 50 years, it has so far not been possible to elucidate the mechanism of action and to use it for improvement. This was not possible because the basis of the mechanism of action of CP, which was found by lucky coincidence, is apoptosis, the discovery of which was honored with the Nobel Prize only in 2002. Another reason was that results from cell culture experiments were used to elucidate the mechanism of action, ignoring the fact that in vivo metabolism differs from in vitro conditions. In vitro, toxic acrolein is formed during the formation of the cytotoxic metabolite phosphoreamidemustard (PAM), whereas in vivo proapoptotic hydroxypropanal (HPA) is formed. The CP metabolites formed in sequence 4-hydroxycyclophosphamide (OHCP) are the main cause of toxicity, aldophosphamide (ALDO) is the pharmacologically active metabolite and HPA amplifies the cytotoxic apoptosis initiated by DNA alkylation by PAM. It is shown that toxicity is drastically reduced but anti-tumor activity strongly increased by the formation of ALDO bypassing OHCP. Furthermore, it is shown that the anti-tumor activity against advanced solid P388 tumors that grow on CD2F1 mice is increased by orders of magnitude if DNA damage caused by a modified PAM is poorly repairable. Motivation: Calculating the magnitude of treatment effects or of differences between two groups is a common task in quantitative science. Standard effect size measures based on differences, such as the commonly used Cohen's, fail to capture the treatment-related effects on the data if the effects were not reflected by the central tendency. The present work aims at (i) developing a non-parametric alternative to Cohen’s d, which (ii) circumvents some of its numerical limitations and (iii) involves obvious changes in the data that do not affect the group means and are therefore not captured by Cohen’s d.
Results: We propose "Impact” as a novel non-parametric measure of effect size obtained as the sum of two separate components and includes (i) a difference-based effect size measure implemented as the change in the central tendency of the group-specific data normalized to pooled variability and (ii) a data distribution shape-based effect size measure implemented as the difference in probability density of the group-specific data. Results obtained on artificial and empirical data showed that “Impact”is superior to Cohen's d by its additional second component in detecting clearly visible effects not reflected in central tendencies. The proposed effect size measure is invariant to the scaling of the data, reflects changes in the central tendency in cases where differences in the shape of probability distributions between subgroups are negligible, but captures changes in probability distributions as effects and is numerically stable even if the variances of the data set or its subgroups disappear.
Conclusions: The proposed effect size measure shares the ability to observe such an effect with machine learning algorithms. Therefore, the proposed effect size measure is particularly well suited for data science and artificial intelligence-based knowledge discovery from big and heterogeneous data.
The subfamily Bromelioideae is one of the most diverse groups among the neotropical Bromeliaceae. Previously, key innovations have been identified which account for the extraordinary radiation and species richness of this subfamily, especially in the so-called core Bromelioideae. However, in order to extend our understanding of the evolutionary mechanisms, the genomic mechanisms (e.g. polyploidy, dysploidy) that potentially underlie this accelerated speciation also need to be tested. Here, using PI and DAPI staining and flow cytometry we estimated genome size and GC content of 231 plants covering 30 genera and 165 species and combined it with published data. The evolutionary and ecological significance of all three genomic characters was tested within a previously generated dated phylogenetic framework using ancestral state reconstructions, comparative phylogenetic methods, and multiple regressions with climatic variables. The absolute genome size (2C) of Bromelioideae varied between 0.59 and 4.11 pg, and the GC content ranged between 36.73 and 41.43%. The monoploid genome sizes (Cx) differed significantly between core and early diverging lineages. The occurrence of dysploidy and polyploidy was, with few exceptions, limited to the phylogenetically isolated early diverging tank-less lineages. For Cx and GC content Ornstein–Uhlenbeck models outperformed the Brownian motion models suggesting adaptive potential linked to the temperature conditions. 2C-values revealed different rates of evolution in core and early diverging lineages also related to climatic conditions. Our results suggest that polyploidy is not associated with higher net diversification and fast radiation in core bromelioids. On the other hand, although coupled with higher extinction rates, dysploidy, polyploidy, and resulting genomic reorganizations might have played a role in the survival of the early diverging bromelioids in hot and arid environments.
Cognitive flexibility – the ability to adjust one’s behavior to changing environmental demands – is crucial for controlled behavior. However, the term ‘cognitive flexibility’ is used heterogeneously, and associations between cognitive flexibility and other facets of flexible behavior have only rarely been studied systematically. To resolve some of these conceptual uncertainties, we directly compared cognitive flexibility (cue-instructed switching between two affectively neutral tasks), affective flexibility (switching between a neutral and an affective task using emotional stimuli), and feedback-based flexibility (non-cued, feedback-dependent switching between two neutral tasks). Three experimental paradigms were established that share as many procedural features (in terms of stimuli and/or task rules) as possible and administered in a pre-registered study plan (N = 100). Correlation analyses revealed significant associations between the efficiency of cognitive and affective task switching (response time switch costs). Feedback-based flexibility (measured as mean number of errors after rule reversals) did not correlate with task switching efficiency in the other paradigms, but selectively with the effectiveness of affective switching (error rate costs when switching from neutral to emotion task). While preregistered confirmatory factor analysis (CFA) provided no clear evidence for a shared factor underlying the efficiency of switching in all three domains of flexibility, an exploratory CFA suggested commonalities regarding switching effectiveness (accuracy-based switch costs). We propose shared mechanisms controlling the efficiency of cue-dependent task switching across domains, while the relationship to feedback-based flexibility may depend on mechanisms controlling switching effectiveness. Our results call for a more stringent conceptual differentiation between different variants of psychological flexibility.
Aerobic and resistance exercise acutely increase cognitive performance (CP). High-intensity functional training (HIFT) combines the characteristics of both regimes but its effect on CP is unclear. Thirty-five healthy individuals (26.7 ± 3.6 years, 18 females) were randomly allocated to three groups. The first (HIFT) performed a functional whole-body workout at maximal effort and in circuit format, while a second walked at 60% of the heart rate reserve (WALK). The third group remained physically inactive reading a book (CON). Before and after the 15-min intervention period, CP was assessed with the Stroop Test, Trail Making Test and Digit Span Test. Repeated-measures ANOVAs and post-hoc 95% confidence intervals (95% CI) were used to detect time/group differences. A significant group*time interaction was found for the backwards condition of the Digit Span Test (p = 0.04) and according to the 95% CI, HIFT was superior to WALK and CON. Analysis of the sum score of the Digit Span Test and the incongruent condition of the Stroop Test, furthermore, revealed main effects for time (p < 0.05) with HIFT being the only intervention improving CP. No differences were found for the Trail Making Test (p > 0.05). In conclusion, HIFT represents an appropriate method to acutely improve working memory, potentially being superior to moderate aerobic-type exercise.
Aufgrund der §§ 20, 44 Abs. 1 Nr. 1 des Hessischen Hochschulgesetzes in der Fassung vom 14. Dezember 2009, zuletzt geändert durch Gesetz vom 27. Mai 2013, hat der Fachbereichsrat des Fachbereichs Physik der Johann Wolfgang Goethe-Universität Frankfurt am Main am 20. Mai 2020 die folgende Ordnung für den Masterstudiengang Physik beschlossen. This afterword addresses the complex temporal and global dynamics of the coronavirus pandemic. After considering some of the new social rhythms that have emerged in the wake of Covid-19 around the world, it turns to the role of collective memory before, during and after corona. The aim is to provide a basic grid for how the Covid-19 pandemic could be addressed using memory studies expertise and concepts such as premediation, memorability, memory (ab)use, national memory, colonial memory, racial stereotypes, the digital archive, generational memory, or Anthropocene time.
Das Ziel dieser Arbeit ist die realitätsgetreue Entwicklung eines interaktiven 3D-Stadtmodells, welches auf den ÖPNV zugeschnitten ist. Dabei soll das Programm anhand von Benutzereingaben und mit Hilfe einer Datenquelle, automatisch eine dreidimensionale Visualisierung der Gebäude erzeugen und den lokalen ÖPNV mitintegrieren. Als Beispiel der Ausarbeitung diente das ÖPNV-Netz der Stadt Frankfurt. Hierbei wurde auf die Problematik der Erhebung von Geoinformationen und der Verarbeitung von solchen komplexen Daten eingegangen. Es wurde ermittelt, welche Nutzergruppen einen Mehrwert durch eine derartige 3D Visualisierung haben und welche neuen Erweiterungs- und Nutzungspotenziale das Modell bietet.
Dem Leser soll insbesondere ein Einblick in die Generierung von interaktiven 3D-Modellen aus reinen Rohdaten verschafft werden. Dazu wurde als Entwicklungsumgebung die Spiele-Engine Unity eingesetzt, welche sich als sehr fähiges und modernes Entwicklungswerkzeug bei der Erstellung von funktionalen 3D-Visualisierungen herausgestellt hat. Als Datenquelle wurde das OpenStreetMap Projekt benutzt und im Rahmen dieser Arbeit behandelt. Anschließend wurde zur Evaluation, das Modell verschiedenen Nutzern bereitgestellt und anhand eines Fragebogens evaluiert.
Während meiner Promotion habe ich zwei Projekte unter der Aufsicht von Dr. Misha Kudryashev durchgeführt. Im ersten Projekt habe ich die Strukturen des Ryanodinrezeptors 1 (RyR1) in Apo- und Ryanodin-Bindungszuständen in der nativen Membran durch Tomographie und Subtomogramm-Mittelung bei 12,6 bzw. 17,5 Å bestimmt. Im Vergleich zur Struktur von gereinigtem RyR1 unter Verwendung der Einzelpartikel-Kryo-Elektronenmikroskopie (Cryo-EM) können zusätzliche Dichten in der cytoplasmatischen Domäne und der sarkoplasmatischen Retikulum (SR)-Membran bzw. im SR-Lumen beobachtet werden. Die Auflösung der Struktur von RyR1 im Apo-Zustand wurde von den Kollegen in meinem Labor mithilfe der Hybridmethode auf 9,5 Å verbessert. Diese Arbeit hat unser Verständnis für die Mechanismen von RyR1 in nativen Membranen erweitert. Im zweiten Projekt habe ich die Struktur des Proteins SdeC der SidE-Familie durch Einzelpartikel-Kryo-EM bei 4,6 Å bestimmt. Die Kristallstruktur des C-Terminus von SdeA wurde von meinem Forschungspartner Dr. Mohit Misra gelöst. Durch Überlagerung einer gemeinsamen Helix dieser beiden Strukturen konnten wir ein kombiniertes Modell erstellen und ein allgemeines Verständnis der Proteine der SidE-Familie erhalten.
Objective: To evaluate if 3 peptides derived from the cartilage oligomeric matrix protein (COMP), which wounded zones of cartilage secrete into synovial fluid, possess biological activity and might therefore be involved in the regulation of specific aspects of joint regeneration.
Methods: The 3 peptides were produced by chemical synthesis and then tested in vitro for known functions of the COMP C-terminal domain from which they derive, and which are involved in osteoarthritis: transforming growth factor-β (TGF-β) signaling, vascular homeostasis, and inflammation. Results. None of the peptides affected the gene expression of COMP in osteochondral progenitor cells (P > 0.05). We observed no effects on the vascularization potential of endothelial cells (P > 0.05). In cultured synovium explants, no differences on the expression of catabolic enzymes or proinflammatory cytokines were found when peptides were added (P > 0.05).
Discussion and conclusions: The 3 peptides tested do not regulate TGF-β signaling, angiogenesis and vascular tube formation, or synovial inflammation in vitro and therefore most likely do not play a major role in the disease process.
Background: The number of implantable cardioverter defibrillator (ICD) infections is increasing due to an increased number of ICD implants, higher-risk patients, and more frequent replacement procedures, which carry a higher risk of infection. Reducing the morbidity, mortality, and cost of ICD-related infections requires an understanding of the current rate of this complication and its predictors.
Methods: The Shock Implant Evaluation Trial (SIMPLE) trial randomized 2500 ICD recipients to defibrillation testing or not. Over an average of 3.1 years, patients were seen every 6 months and examined for evidence of ICD infection, which was defined as requiring device removal and/or intravenous antibiotics.
Results: Within 24 months, 21 patients (0.8%) developed infection. Fourteen patients (67%) with infection presented within 30 days, 20 patients by 12 months, and only 1 patient beyond 12 months. Univariate analysis demonstrated that patients with primary electrical disorders (3 patients, P = 0.009) and those with a secondary prevention indication (13 patients, P = 0.0009) were more likely to develop infection. Among the 2.2% of patients who developed an ICD wound hematoma, 10.4% developed an infection. Among the 8.3% of patients requiring an ICD reintervention, 1.9% developed an infection.
Conclusions: This cohort of ICD recipients at high-volume centres have a low risk of device-related infection. However; strategies to reduce wound hematoma and the need for ICD reintervention could further reduce the rate of infection.
This article reports an investigation of how inhibition contributes to fluid reasoning when it is decomposed into the reasoning ability, item-position, and speed components to control for possible method effects. Working memory was also taken into consideration. A sample of 223 university students completed a fluid reasoning scale, two tasks tapping prepotent response inhibition, and two working memory tasks. Fixed-links modeling was used to separate the effect of reasoning ability from the effects of item-position and speed. The goodness-of-fit results confirmed the necessity to consider the reasoning ability, item-position, and speed components simultaneously. Prepotent response inhibition was only associated with reasoning ability. This association disappeared when working memory served as a mediator. Taken together, these results reflect the inhomogeneity of what is tapped by the fluid reasoning scale on one hand and, on the other, suggest inhibition as an important component of working memory.

On the basis of the economic theory of network effects, this article provides a novel explanation of the so-called patent paradox, i.e. the question why the propensity to patent is so strong when the expected average value of most patents is low. It demonstrates that the patent system of a country resembles a telephone network or a social media platform. Patents are perceived as nodes in a virtual network that, as a whole, exhibits network effects. It is explained why patents are not independent of other patents but that they complement each other in several ways both within and beyond markets and fields of technology, and that patents thus create synchronization value over and above individual interests of patent holders in exclusivity. As a consequence, the more patents there are, the more valuable it is to also seek patents, and vice versa. Since patents thus display increasing returns to adoption, the willingness to pay for the next patent slopes upwards. This explains why, after a phase of early instability and a certain tipping point, many countries’ patent systems expanded quickly and eventually became a rigid standard (“lock-in”). The concluding section raises the question what regulatory measures are suitable to effectively address the ensuing anticommons effects.
The long-standing battle between economic nationalism and globalism has again taken center stage in geopolitics. This article applies this dichotomy to the law and policy of international intellectual property (IP). Most commentators see IP as a prime example of globalization. The article challenges this view on several levels. In a nutshell, it claims that economic nationalist concerns about domestic industries and economic development lie at the heart of the global IP system. To support this argument, the article summarizes and categorizes IP policies adopted by selected European countries, the European Union, and the U.S. Section I presents three types of inbound IP policies that aim to foster local economic development and innovation. Section II adds three versions of outbound IP policies that, in contrast, target foreign countries and markets. Concluding section III traces a dialectic virtuous circle of economic nationalist motives leading to global legal structures and identifies the function and legal structure of IP as the reason for the resilience and even dominance of economic nationalist motives in international IP politics. IP concerns exclusive private rights that are territorially limited creatures of (supra-)national statutes. These legal structures make up the economic nationalist DNA of IP.
Der Beitrag erläutert aus rechtswissenschaftlicher Sicht, welche Begriffe, Perspektiven und Methoden zur Beantwortung folgender Frage geeignet sind: Wie lässt sich erklären, dass das gerade einmal ca. 200 Jahre alte Rechtsgebiet des „geistigen Eigentums“, dessen Legitimität stets heftig umstritten war, praktisch weltweite Anerkennung in nationalen, supranationalen und völkerrechtlichen Rechtsquellen finden konnte und die globalen Handels- und Kommunikationsströme auch tatsächlich wirksam reguliert? Oder anders gewendet: Wie kann ein schwacher Geltungsanspruch eine faktisch wirkmächtige Ordnung hervorbringen? Nach kritisch-diskurstheoretischen Ansätzen bildet der Streit um Rechtfertigungen den zentralen Treiber normativer Ordnungen. Der Aufsatz zeigt jedoch, dass im Hinblick auf das „geistige Eigentum“ normexterne Bedingungen wie technischer Fortschritt und der Wandel der allgemeinen Wirtschaftsordnung von entscheidender Bedeutung waren und der Frage nach den Gründen z.T. logisch vorausliegen. Schließlich wird gezeigt, dass sozialontologische Analysen besonders gut geeignet erscheinen, strukturelle Normativität/Macht zu adressieren, die unterhalb expliziter Rechtfertigungen bzw. im „Hintergrund“ angesiedelt ist, so wie Sprache generell die logische Voraussetzung für ein rechtfertigendes Rede-und-Antwort-Stehen bildet.
Der Beitrag stellt zunächst dar, dass und warum dem geltenden UWG ein schlüssiger innerer Zusammenhang fehlt, und zwar sowohl im Hinblick auf den Prüfungsaufbau für Ansprüche als auch im Hinblick auf die im Gesetz zum Tragen kommenden materiellen Systematisierungskriterien. Sodann wird ein Vorschlag für einen alternativen Gesetzesaufbau unterbreitet, der am unverändert richtigen Gedanken festhält, wonach Wettbewerbsverhalten in seinen komplexen Auswirkungen auf sämtliche Marktteilnehmer ganzheitlich zu erfassen und zu regulieren ist, während die Orientierung an Gruppen von Marktteilnehmern zwangsläufig zu Abgrenzungsproblemen führt. Folglich bilden im unterbreiteten Vorschlag nicht die Schutzsubjekte des UWG, sondern der Unwertgehalt der geschäftlichen Handlung (Irreführung, Aggressivität, sonstige Wettbewerbsverfälschung) das primäre Systematisierungskriterium. Übergeordnetes Ziel des Vorschlags ist es, die methodengerechte Anwendung des geschriebenen Rechts zu erleichtern und auf diese Weise gesetzesferne Eigenrationalitäten der Praxis einzuhegen.
Der Beitrag nimmt zu den Entscheidungen des BGH v. 21.2.2019 in Sachen HHole (for Mannheim) u.a., wonach die Vernichtung eines urheberrechtlich geschützten Werks eine „andere Beeinträchtigung“ im Sinne des § 14 UrhG darstellt, kritisch Stellung. Hinterfragt wird insbesondere, worauf sich Entstellungen oder Beeinträchtigungen gem. § 14 UrhG beziehen, was also unter einem „Werk“ gem. § 14 UrhG zu verstehen ist.
Die rechtshistorische Forschung konzentriert sich auf die Herausbildung spezifisch juristischer Figuren wie das frühneuzeitliche Privileg oder das spätere „geistige Eigentum“. Die Geschichte des urheberrechtlichen Werkbegriffs – immerhin der Gegenstand der „Werkherrschaft“ des Urhebers – hat demgegenüber verhältnismäßig wenig Aufmerksamkeit erfahren. Dabei ist letztlich unstreitig, dass auch die Vorstellung eines von seinen Verkörperungen unabhängigen und daher abstrakt-immateriellen, eigentumsfähigen Werks keine gegebene Wirklichkeit darstellt, sondern erst seit dem Ende des 18. Jahrhunderts etabliert ist. Der Beitrag historisiert das Urheberrecht ausgehend von seiner – so die Hypothese – vorgestellten Wirklichkeit. Betrachtet werden drei Vorbedingungen, die das juristische Reden und Denken in Kategorien eines abstrakt-unkörperlichen, eigentumsfähigen Werks und damit zugleich das „geistige Eigentum“ an diesem Werk möglich gemacht haben: Reproduktionstechnologien, das Ideal des genialen Werks und die dezentral-anonyme Marktwirtschaft.
Das Netzwerkdurchsetzungsgesetz (NetzDG) ist einseitig auf die Löschung rechtswidriger Inhalte ausgerichtet. Vorzugswürdig erscheint demgegenüber ein ganzheitlicher Regulierungsansatz, der darauf abzielt, dass Anbieter großer Universalplattformen gleichermaßen rechtswidrige Inhalte löschen und rechtmäßige Kommunikation nicht willkürlich beeinträchtigen. Dazu wird hier ein konkreter Entwurf zur Änderung des NetzDG nebst Begründung vorgelegt.
Using a structural life-cycle model, we quantify the long-term impact of school closures during the Corona crisis on children affected at different ages and coming from households with different parental characteristics. In the model, public investment through schooling is combined with parental time and resource investments in the production of child human capital at different stages in the children's development process. We quantitatively characterize both the long-term earnings consequences on children from a Covid-19 induced loss of schooling, as well as the associated welfare losses. Due to self-productivity in the human capital production function, skill attainment at a younger stage of the life cycle raises skill attainment at later stages, and thus younger children are hurt more by the school closures than older children. We find that parental reactions reduce the negative impact of the school closures, but do not fully offset it. The negative impact of the crisis on children's welfare is especially severe for those with parents with low educational attainment and low assets. The school closures themselves are primarily responsible for the negative impact of the Covid-19 shock on the long-run welfare of the children, with the pandemic-induced income shock to parents playing a secondary role.
Les données de l’effet d’affaiblissement de l’obviation (ou encore référence disjointe du subjonctif) dans la littérature linguistique sont soit fondées sur l’intuition des auteurs soit issues de publications d’autres chercheurs, bien que cet effet soit connu depuis longtemps et bien qu’il fasse partie de plusieurs modélisations théoriques. En conséquence, aucune réponse de nature empirique/expérimentale a été offerte à la question « A quel point les affirmations portant sur l'effet d'affaiblissement de l'obviation sont-elles solides ? ». Cet article se fixe en conséquence un double objectif : (i) mettre en oeuvre un jugement de grammaticalité afin de répondre expérimentalement à la question précédente, et (ii) proposer une analyse syntaxique provisoire des résultats obtenus. Ici, nous étudions six facteurs qui – selon Ruwet (1984/1991) – déclenchent l’affaiblissement. Les résultats montrent que seul un facteur (la coordination) déclenche l’affaiblissement. En conséquence, l’appareil théorique permettant de modéliser l’affaiblissement peut être réduit radicalement, et nous proposons des pistes de modélisation syntaxique n’intégrant que le facteur de coordination
In this explorative study, we investigate how sequences of behaviour are related to success or failure in complex problem‐solving (CPS). To this end, we analysed log data from two different tasks of the problem‐solving assessment of the Programme for International Student Assessment 2012 study (n = 30,098 students). We first coded every interaction of students as (initial or repeated) exploration, (initial or repeated) goal‐directed behaviour, or resetting the task. We then split the data according to task successes and failures. We used full‐path sequence analysis to identify groups of students with similar behavioural patterns in the respective tasks. Double‐checking and minimalistic behaviour was associated with success in CPS, while guessing and exploring task‐irrelevant content was associated with failure. Our findings held for both tasks investigated, from two different CPS measurement frameworks. We thus gained detailed insight into the behavioural processes that are related to success and failure in CPS.

Chronische pulmonale Infektionen mit Pseudomonas aeruginosa (PA) betreffen die überwiegende Mehrheit der erwachsenen Mukoviszidose (Cystische Fibrose, CF) Patienten.
Diese Infektionen führen gesichert zu einer Abnahme der Lungenfunktion und Zunahme der Mortalität der Patienten. Atemwegsviren stehen im Verdacht pulmonale Exazerbationen bei CF-Patienten auszulösen. Unklar ist jedoch, welchen Einfluss eine chronische Infektion mit PA auf die Anfälligkeit und Reaktion des Atemwegsepithels auf virale Infektionen hat.
Das Ziel dieser Arbeit war es daher, die Interaktionen zwischen PA, humanen Rhinoviren (HRV) und primären bronchialen Epithelzellen zu untersuchen. Hierfür wurden Zellen von jeweils drei Patienten mit CF und mit Lungenemphysem aus Lungenexplantaten isoliert und in einem speziellen Air- Liquid-Interface Zellkulturmodell zu einem mukoziliär differenzierten mehrreihigen Flimmerepithel kultiviert. Chronische Infektionen wurden mit klinischen PA Isolaten für einen Gesamtzeitraum von 16 Tagen durchgeführt. Anschließend wurden die Zellen mit HRV infiziert. Schlüsselzytokine, Interferone und virale RNA wurden mittels Cytometric bead array, ELISA und qPCR bestimmt.
Rein virale Infektionen mit HRV führten zu einem Anstieg von IL-1, -6, -8, TNF- α, IP10 und IFN-b, IFN-l1 sowie ISGs und in ähnlichem Ausmaß konnte dies auch bei Coinfektionen mit einem mukoiden PA-Isolat beobachtet werden. Coinfektionen mit einem nicht-mukoiden PA-Isolat führten im Vergleich zu rein viralen Infektionen zu vermehrter Expression von IL-1β und IL-6 mRNA. Während es unter diesen Bedingungen auch auf Proteinebene zu einem Anstieg der IL-1β Konzentration kam, lag die Konzentration von freiem IL-6 Protein in nahezu allen Proben unter der Nachweisgrenze. Zellkulturmedium aus Coinfektionen mit diesem nicht-mukoiden PA-Isolat führten zudem zu einem Abbau oder einer Bindung von extern zugegebenen rekombinantem IL-6.
IL-8, IP-10, TNF-α Protein und mRNA von IFN-β, -λ1 und ISGs, sowie die Viruslast waren vergleichbar zwischen rein viralen Infektionen und bakteriell- viralen Coinfektionen. Ebenfalls keine Unterschiede wurden zwischen Zellen von Emphysem und CF-Spendern gefunden. Insgesamt zeigen diese Daten, dass eine PA-Infektion die Antwort differenzierter bronchialer Epithelzellen auf eine Virusinfektion verändern kann. Die hierdurch veränderte Immunantwort und möglicherweise eingeschränkten epithelialen Reparaturmechanismen könnten eine Ursache aggravierter viraler Infektionen in P. aeruginosa-infizierten Atemwegen darstellen.
Ein besseres Verständnis der Interaktionen zwischen chronisch-bakteriellen und viralen Atemwegsinfektionen könnte potenziell die Behandlung virus-induzierter Exazerbationen bei PA-infizierten CF-Patienten verbessern.
Currently, due to the misuse of antibiotics, we are facing a major public health problem. The resistance to antibiotics of certain bacterial strains makes the treatment of infections very complex.
In this context, the present thesis project concerns the study of a bacterial efflux complex capable of transporting antibiotics from the cytoplasm to the outside of the cell. This complex is composed of an inner-membrane Major Facilitator Superfamily (MFS) transporter (EmrB, E. coli multidrug resistance), a channel of the outer membrane TolC (Tolerance to Colicin E1) and a periplasmic adapter (EmrA, E. coli multidrug resistance). Unlike RND-type efflux systems (such as AcrAB-TolC), little is known about the MFS-type EmrAB-TolC system. It is therefore important to study the entire complex on a structural and functional level, to analyse the marked differences between these two types of transport systems. The goal of my thesis project was to study at least one EmrAB-TolC complex from a structural point of view. For my studies the aim was to isolate the complex directly from bacteria overexpressing the three protein partners. In a first step, 15 homologous EmrAB-TolC systems were identified and their corresponding genes amplified from genomic DNA of different Gram-negative bacteria. Among the genes of the 15 systems, the genes coding for the E. coli and V. cholerae systems were further studied. The expression vectors encoded fluorescent markers for the monitoring of the expression levels of different proteins and for studying the formation of complexes. In a first step, the different protein expression levels (EmrB-mRFP1 and EmrA-sfGFP) were studied for several expression strains of E. coli by measuring the red and green fluorescence levels and by Western blot (anti-His, Myc, and Strep for EmrB, EmrA, and TolC). The E. coli strain C41(DE3) was best suited for co-expression of EmrAB-TolC. In a second step, the FSEC (Fluorescence detection Size Exclusion Chromatography) methodology was used to identify a complex suitable for structural study. Thus this method enabled the observation that the EmrAB-TolC complex of E. coli was produced in higher amount than that of V. cholerae. The final co-purification protocol consists in perfoming a gentle lysis of the bacteria using lysozyme, then after solubilization with DDM, the purification is started by a Ni2+-NTA affinity chromatography step followed by a size exclusion chromatography step. Finally, the fractions containing the three protein partners are used for the detergent-exchange by amphipol A8-35 before the structural study by electron microscopy. Negative stain EM-micrographs displayed elongated objects with a length of 33 nm in side view. An average image of EmrAB-TolC shows similarities to that of the AcrAB-TolC complex observed under similar conditions. Similarities included the characteristic densities of TolC. Whereas differences were found in the lower part of EmrAB which is thinner than the lower part of AcrAB. The densities visible above the amphipol-ring correspond to EmrA, which displays a channel-like structure as in AcrA. The channel however seems to extend further towards the amphipol belt. Since EmrB does not have an extended periplasmic domain as the RND proteins have, these densities are therefore solely assigned to EmrA. EmrA, on the other side, contacts TolC akin to the interaction of AcrA/MexA to their cognate outer membrane channels (TolC/OprM) in a ‘tip-to-tip’ fashion.
Spinocerebellar ataxia type 2 (SCA2) is caused by polyglutamine expansion in Ataxin-2 (ATXN2). This factor binds RNA/proteins to modify metabolism after stress, and to control calcium (Ca2+) homeostasis after stimuli. Cerebellar ataxias and corticospinal motor neuron degeneration are determined by gain/loss in ATXN2 function, so we aimed to identify key molecules in this atrophic process, as potential disease progression markers. Our Atxn2-CAG100-Knock-In mouse faithfully models features observed in patients at pre-onset, early and terminal stages. Here, its cerebellar global RNA profiling revealed downregulation of signaling cascades to precede motor deficits. Validation work at mRNA/protein level defined alterations that were independent of constant physiological ATXN2 functions, but specific for RNA/aggregation toxicity, and progressive across the short lifespan. The earliest changes were detected at three months among Ca2+ channels/transporters (Itpr1, Ryr3, Atp2a2, Atp2a3, Trpc3), IP3 metabolism (Plcg1, Inpp5a, Itpka), and Ca2+-Calmodulin dependent kinases (Camk2a, Camk4). CaMKIV–Sam68 control over alternative splicing of Nrxn1, an adhesion component of glutamatergic synapses between granule and Purkinje neurons, was found to be affected. Systematic screening of pre/post-synapse components, with dendrite morphology assessment, suggested early impairment of CamKIIα abundance together with the weakening of parallel fiber connectivity. These data reveal molecular changes due to ATXN2 pathology, primarily impacting excitability and communication.
Lichen-forming fungi are symbiotic organisms that synthesize unique natural products with potential for new drug leads. Here, we explored the pharmacological activity of six lichen extracts (Evernia prunastri, Pseudevernia furfuracea, Umbilicaria pustulata, Umbilicaria crustulosa, Flavoparmelia caperata, Platismatia glauca) in the context of cancer and inflammation using a comprehensive set of 11 functional and biochemical in vitro screening assays. We assayed intracellular Ca2+ levels and cell migration. For cancer, we measured tumor cell proliferation, cell cycle distribution and apoptosis, as well as the angiogenesis-associated proliferation of endothelial cells (ECs). Targeting inflammation, we assayed leukocyte adhesion onto ECs, EC adhesion molecule expression, as well as nitric oxide production and prostaglandin (PG)E2 synthesis in leukocytes. Remarkably, none of the lichen extracts showed any detrimental influence on the viability of ECs. We showed for the first time that extracts of F. caperata induce Ca2+ signaling. Furthermore, extracts from E. prunastri, P. furfuracea, F. caperata, and P. glauca reduced cell migration. Interestingly, F. caperata extracts strongly decreased tumor cell survival. The proliferation of ECs was significantly reduced by E. prunastri, P. furfuracea, and F. caperata extracts. The extracts did not inhibit the activity of inflammatory processes in ECs. However, the pro-inflammatory activation of leukocytes was inhibited by extracts from E. prunastri, P. furfuracea, F. caperata, and P. glauca. After revealing the potential biological activities of lichen extracts by an array of screening tests, a correlation analysis was performed to evaluate particular roles of abundant lichen secondary metabolites, such as atranorin, physodic acid, and protocetraric acid as well as usnic acid in various combinations. Overall, some of the lichen extracts tested in this study exhibit significant pharmacological activity in the context of inflammation and/or cancer, indicating that the group lichen-forming fungi includes promising members for further testing.
Nitro fatty acids (NFAs) are endogenously generated lipid mediators deriving from reactions of unsaturated electrophilic fatty acids with reactive nitrogen species. Furthermore, Mediterranean diets can be a source of NFA. These highly electrophilic fatty acids can undergo Michael addition reaction with cysteine residues, leading to post-translational modifications (PTM) of selected regulatory proteins. Such modifications are capable of changing target protein function during cell signaling or in biosynthetic pathways. NFA target proteins include the peroxisome proliferator-activated receptor γ (PPAR-γ), the pro-inflammatory and tumorigenic nuclear factor-κB (NF-κB) signaling pathway, the pro-inflammatory 5-lipoxygenases (5-LO) biosynthesis pathway as well as soluble epoxide hydrolase (sEH), which is essentially involved in the regulation of vascular tone. In several animal models of inflammation and cancer, the therapeutic efficacy of well-tolerated NFA has been demonstrated. This has already led to clinical phase II studies investigating possible therapeutic effects of NFA in subjects with pulmonary arterial hypertension. Albeit Michael acceptors feature a broad spectrum of bioactivity, they have for a rather long time been avoided as drug candidates owing to their presumed unselective reactivity and toxicity. However, targeted covalent modification of regulatory proteins by Michael acceptors became recognized as a promising approach to drug discovery with the recent FDA approvals of the cancer therapeutics, afatanib (2013), ibrutinib (2013), and osimertinib (2015). Furthermore, the Michael acceptor, neratinib, a dual inhibitor of the human epidermal growth factor receptor 2 and epidermal growth factor receptor, was recently approved by the FDA (2017) and by the EMA (2018) for the treatment of breast cancer. Finally, a number of further Michael acceptor drug candidates are currently under clinical investigation for pharmacotherapy of inflammation and cancer. In this review, we focus on the pharmacology of NFA and other Michael acceptor drugs, summarizing their potential as an emerging class of future antiphlogistics and adjuvant in tumor therapeutics.
Post-exercise hypotension (PEH) is the phenomenon of lowered blood pressure after a single bout of exercise. Only a fraction of people develops PEH but its occurrence correlates well with long-term effects of sports on blood pressure. Therefore, PEH has been suggested as a suitable predictor for the effectivity of exercise as therapy in hypertension. Local vascular bioactive lipids might play a potential role in this context. We performed a cross-over clinical pilot study with 18 healthy volunteers to investigate the occurrence of PEH after a single short-term endurance exercise. Furthermore, we investigated the plasma lipid profile with focus on arachidonic acid (AA)-derived metabolites as potential biomarkers of PEH. A single bout of ergometer cycling induced a significant PEH in healthy volunteers with the expected high inter-individual variability. Targeted lipid spectrum analysis revealed significant upregulation of several lipids in the direct post-exercise phase. Among these changes, only 15- hydroxyeicosatetranoic acid (HETE) correlated significantly with the extent of PEH but in an AA-independent manner, suggesting that 15-HETE might act as specific PEH-marker. Our data indicate that specific lipid modulation might facilitate the identification of patients who will benefit from exercise activity in hypertension therapy. However, larger trials including hypertonic patients are necessary to verify the clinical value of this hypothesis.
The risk of increasing dengue (DEN) and chikungunya (CHIK) epidemics impacts 240 million people, health systems, and the economy in the Hindu Kush Himalayan (HKH) region. The aim of this systematic review is to monitor trends in the distribution and spread of DEN/CHIK over time and geographically for future reliable vector and disease control in the HKH region. We conducted a systematic review of the literature on the spatiotemporal distribution of DEN/CHIK in HKH published up to 23 January 2020, following Preferred Reporting Items for Systematic Reviews and Meta-Analysis (PRISMA) guidelines. In total, we found 61 articles that focused on the spatial and temporal distribution of 72,715 DEN and 2334 CHIK cases in the HKH region from 1951 to 2020. DEN incidence occurs in seven HKH countries, i.e., India, Nepal, Bhutan, Pakistan, Bangladesh, Afghanistan, and Myanmar, and CHIK occurs in four HKH countries, i.e., India, Nepal, Bhutan, and Myanmar, out of eight HKH countries. DEN is highly seasonal and starts with the onset of the monsoon (July in India and June in Nepal) and with the onset of spring (May in Bhutan) and peaks in the postmonsoon season (September to November). This current trend of increasing numbers of both diseases in many countries of the HKH region requires coordination of response efforts to prevent and control the future expansion of those vector-borne diseases to nonendemic areas, across national borders.
Background: While swallowing disorders are frequent sequela following posterior fossa tumor (PFT) surgery in children, data on dysphagia frequency, severity, and outcome in adults are lacking. The aim of this study was to investigate dysphagia before and after surgical removal of PFT. Additionally, we tried to identify clinical predictors for postsurgical swallowing disorders. Furthermore, this study explored the three-month outcome of dysphagic patients.
Methods: In a cohort of patients undergoing PFT surgery, dysphagia was prospectively assessed pre- and postoperatively using fiberoptic endoscopic evaluation of swallowing. Patients with severe dysphagia at discharge were re-evaluated after three months. Additionally, clinical and imaging data were collected to identify predictors for post-surgical dysphagia. Results: We included 26 patients of whom 15 had pre-operative swallowing disorders. After surgery, worsening of pre-existing dysphagia could be noticed in 7 patients whereas improvement was observed in 2 and full recovery in 3 subjects. New-onset dysphagia after surgery occurred in a minority of 3 cases. Postoperatively, 47% of dysphagic patients required nasogastric tube feeding. Re-evaluation after three months of follow-up revealed that all dysphagic patients had returned to full oral intake.
Conclusion: Dysphagia is a frequent finding in patients with PFT already before surgery. Surgical intervention can infer a deterioration of impaired swallowing function placing affected patients at temporary risk for aspiration. In contrast, surgery can also accomplish beneficial results resulting in both improvement and full recovery. Overall, our findings show the need of early dysphagia assessment to define the safest feeding route for the patient.
Generic tasks for algorithms

Due to its links to computer science (CS), teaching computational thinking (CT) often involves the handling of algorithms in activities, such as their implementation or analysis. Although there already exists a wide variety of different tasks for various learning environments in the area of computer science, there is less material available for CT. In this article, we propose so-called Generic Tasks for algorithms inspired by common programming tasks from CS education. Generic Tasks can be seen as a family of tasks with a common underlying structure, format, and aim, and can serve as best-practice examples. They thus bring many advantages, such as facilitating the process of creating new content and supporting asynchronous teaching formats. The Generic Tasks that we propose were evaluated by 14 experts in the field of Science, Technology, Engineering, and Mathematics (STEM) education. Apart from a general estimation in regard to the meaningfulness of the proposed tasks, the experts also rated which and how strongly six core CT skills are addressed by the tasks. We conclude that, even though the experts consider the tasks to be meaningful, not all CT-related skills can be specifically addressed. It is thus important to define additional tasks for CT that are detached from algorithms and programming.
Background:Aedes aegypti is a potential vector for several arboviruses including dengue and Zika viruses. The species seems to be restricted to subtropical/tropical habitats and has difficulties in establishing permanent populations in southern Europe, probably due to constraints during the winter season. The aim of this study was to systematically analyze the cold tolerance (CT) of Ae. aegypti in its most cold-resistant life stage, the eggs.
Methods: The CT of Ae. aegypti eggs was compared with that of Ae. albopictus which is well established in large parts of Europe. By systematically studying the literature (meta-analysis), we recognized that CT has been rarely tested in Ae. aegypti eggs, but eggs can survive at zero and sub-zero temperatures for certain exposure periods. To overcome potential bias from experimental differences between studies, we then conducted species comparisons using a harmonized high-resolution CT measuring method. From subtropical populations of the same origin, the survival (hatching in %) and emergence of adults of both species were measured after zero and sub-zero temperature exposures for up to 9 days (3 °C, 0 °C and − 2 °C: ≤ 9 days; − 6 °C: ≤ 2 days).
Results: Our data show that Ae. aegypti eggs can survive low and sub-zero temperatures for a short time period similar to or even better than those of Ae. albopictus. Moreover, after short sub-zero exposures of eggs of both species, individuals still developed into viable adults (Ae. aegypti: 3 adults emerged after 6 days at − 2 °C, Ae. albopictus: 1 adult emerged after 1 day at − 6 °C).
Conclusions: Thus, both the literature and the present experimental data indicate that a cold winter may not be the preventing factor for the re-establishment of the dengue vector Ae. aegypti in southern Europe.
Erasmus, christlicher Humanismus und Spiritualität in Spanien und Neu-Spanien (16. Jahrhundert)

Schriften des Erasmus von Rotterdam (1466/69–1536) entfalteten während des 16. Jahrhunderts eine große Wirkung in Spanien. Auf Grundlage der klassischen wie der jüngeren Historiographie widmet sich der Aufsatz diesem religions- und kulturgeschichtlichen Phänomen – mit Seitenblicken auf Luther – in vier Teilen: Nach einer Skizze zu Leben und Werk des christlichen Humanisten behandelt der zweite Teil den Erasmianismus in Spanien von seiner Erfolgsgeschichte in den 1520er Jahren (etwa bei Hof, an den Universitäten und in Übersetzungen) bis zur Verfolgung seiner Anhänger seit den 1530er Jahren durch die Inquisition. Drittens werden neuere Forschungstendenzen diskutiert, die das klassische, von Marcel Bataillon geprägte Bild korrigieren und weiterentwickeln, auch im Hinblick auf das ambivalente Verhältnis von Scholastik und Humanismus. Der letzte Teil widmet sich dem Einfluss des Erasmus in Neuspanien (Mexiko) am Beispiel von (Erz-) Bischöfen und Mönchen sowie von frühkolonialen Fallstudien.
The inflammatory response plays an important role in the pathophysiology of multiple injuries. This study examines the effects of severe trauma and inflammatory response on markers of neuronal damage. A retrospective analysis of prospectively collected data in 445 trauma patients (Injury Severity Score (ISS) ≥ 16) is provided. Levels of neuronal biomarkers (calcium-binding Protein B (S100b), Enolase2 (NSE), glial fibrillary acidic protein (GFAP)) and Interleukins (IL-6, IL-10) in severely injured patients (with polytrauma (PT)) without traumatic brain injury (TBI) or with severe TBI (PT+TBI) and patients with isolated TBI (isTBI) were measured upon arrival until day 5. S100b, NSE, GFAP levels showed a time-dependent decrease in all cohorts. Their expression was higher after multiple injuries (p = 0.038) comparing isTBI. Positive correlation of marker level after concomitant TBI and isTBI (p = 0.001) was noted, while marker expression after PT appears to be independent. Highest levels of IL-6 and -10 were associated to PT und lowest to isTBI (p < 0.001). In all groups pro-inflammatory response (IL-6/-10 ratio) peaked on day 2 and at a lower level on day 4. Severe TBI modulates kinetic profile of inflammatory response by reducing interleukin expression following trauma. Potential markers for neuronal damage have a limited diagnostic value after severe trauma because undifferentiated increase.
Im Rahmen dieser Arbeit wurde ein Reaktionsmikroskop (REMI) nach dem Messprinzip COLTRIMS (Cold Target Recoil Ion Momentum Spectrometry) neu konstruiert und aufgebaut. Die Leistungsfähigkeit des Experimentaufbaus konnte sowohl in diversen Testreihen als auch anschließend unter realen Messbedingungen an der Synchrotronstrahlungsanlage SOLEIL und am endgültigen Bestimmungsort SQS-Instrument (Small Quantum Systems) des Freie-Elektronen-Lasers European XFEL (X-ray free-electron laser) eindrucksvoll unter Beweis gestellt werden.
Mit der Experimentiertechnik COLTRIMS ist es möglich, alle geladenen Fragmente einer Wechselwirkung eines Projektilteilchens mit einem Targetteilchen mittels zweier orts- und zeitauflösender Detektoren nachzuweisen. In einem Vakuumrezipienten wird die als Molekularstrahl präparierte Targetsubstanz inmitten der Hauptkammer zentral mit einem Projektilstrahl (z.B. des XFEL) zum Überlapp gebracht, sodass dort eine Wechselwirkung stattfinden kann. Bei den entstehenden Fragmenten handelt es sich um positiv geladene Ionen sowie negative geladene Elektronen. Elektrische Felder, erzeugt durch eine Spektrometer-Einheit, sowie durch Helmholtz-Spulen erzeugte magnetische Felder ermöglichen es, die geladenen Fragmente in Richtung der Detektoren zu lenken. Die Orts- und Zeitmessung eines einzelnen Teilchens (z.B. eines Ions) findet in Koinzidenz mit den anderen Teilchen (z.B. weiteren Ionen bzw. Elektronen) statt. Mit dieser Messmethode können die Impulsvektoren und Ladungszustände aller geladenen Fragmente in Koinzidenz gemessen werden. Da hierbei die geometrische Anordnung der einzelnen Komponenten für die Leistungsfähigkeit des Experiments eine entscheidende Rolle spielt, mussten bei der Neukonstruktion des COLTRIMS-Apparates für den Einsatz an einem Freie-Elektronen-Laser (FEL) einige Rahmenbedingungen erfüllt werden. Besonders wurden die hohen Vakuumvoraussetzungen an den Experimentaufbau aufgrund der enormen Lichtintensität eines FEL beachtet. Das Zusammenspiel der vielen Einzelkomponenten konnte zunächst in mehreren Testreihen überprüft werden. Unter anderem durch Variation der Vakuumbauteile in Material und Beschaffenheit konnten die zuvor ermittelten Vorgaben schließlich erreicht werden. Das neu konstruierte Target-Präparationssystem zur Erzeugung molekularer Gasstrahlen erlaubt nun den Einsatz von bis zu vier unterschiedlich dimensionierten, differentiell gepumpten Stufen. Zudem wurden hochpräzise Piezo-Aktuatoren verbaut, welche die Bewegung von Blenden im Vakuum erlauben, wodurch eine variable Einstellung des lokalen Targetdrucks ermöglicht wird. Die Anpassung der elektrischen Felder des Spektrometers für ein jeweiliges Experiment wurde mittels Simulationen der Teilchentrajektorien, Teilchenflugzeiten sowie der Detektorauflösung durchgeführt.
Da die in dieser Arbeit besprochenen Messungen und Ergebnisse die Wechselwirkungsprozesse von Röntgenstrahlung bzw. Synchrotronstrahlung mit Materie thematisieren, wird die Erzeugung von Synchrotronstrahlung sowohl in Kreisbeschleunigern als auch in den modernen Freie-Elektronen-Lasern (FEL) erklärt und hergeleitet. Der im Röntgenbereich arbeitende Freie-Elektronen-Laser European XFEL, welcher u.A. als Strahlungsquelle für die hier gezeigten Experimente diente, ist eine von derzeit noch wenigen Anlagen ihrer Art weltweit. Seine Lichtintensität in diesem Wellenlängenbereich liegt bis zu acht Größenordnungen über den bisher verwendeten Anlagen für Synchrotronstrahlung.
Beim ersten Einsatz der neuen Apparatur an der Synchrotronstrahlungsanlage SOLEIL wurde der ultraschnelle Dissoziationsprozess von Chlormethan (CH3Cl) untersucht. Während des Zerfallsprozesses nach Anregung durch Röntgenstrahlung werden hochenergetische Auger-Elektronen emittiert, welche in Koinzidenz mit verschiedenen Molekülfragmenten nachgewiesen wurden. Durch den Zerfallsmechanismus der ultraschnellen Dissoziation wird die Auger-Elektronenemission nach resonanter Molekülanregung während der Dissoziation des Moleküls beschrieben. Die kinetische Energie des Auger-Elektrons ist dabei abhängig von seinem Emissionszeitpunkt. Somit können die gemessenen Auger-Elektronen ein „Standbild“ der zeitlichen Abfolge des Dissoziationsprozesses liefern.
Es wird eine detaillierte Beschreibung der Datenanalyse vorgenommen, welche aus Kalibrationsmessungen und einer Interpretation der Messdaten besteht. Die abschließende Betrachtung besteht in der Darstellung der Elektronenemissionswinkelverteilungen im molekülfesten Koordinatensystem. Die Winkelverteilung der Auger-Elektronen wird am Anfang der Dissoziation vom umgebenden Molekül- potential beeinflusst und zeigt deutliche Strukturen entlang der Bindungsachse. Entfernen sich die Bindungspartner voneinander und das Auger-Elektron wird währenddessen emittiert, so verschwinden diese Strukturen zunehmend und eine Vorzugsemissionsrichtung senkrecht zur Molekülachse wird sichtbar.
Die Analyse der Messdaten zur Untersuchung von Multiphotonen-Ionisation an Sauerstoff-Molekülen am Freie-Elektronen-Laser European XFEL ermöglichte unter anderem die Beobachtung „hohler Moleküle“, also Systemen mit Doppelinnerschalen- Vakanzen. Solche Zustände können vor allem durch die sequentielle Absorption zweier Photonen entstehen, wobei die hierbei nötige Photonendichte nur von FEL- Anlagen bereit gestellt werden kann. Hier konnte das Ziel erreicht werden, erstmalig die Emissionswinkelverteilungen der Photoelektronen von mehrfach ionisierten Sauerstoff-Molekülen (O+/O3+-Aufbruchskanal) als Folge der ablaufenden Mechanismen femtosekundengenau zu beobachten. Hierzu wurde ein vereinfachtes Schema der verschiedenen Zerfallsschritte erstellt und schließlich ermittelt, dass der Zerfall durch eine PAPA-Sequenz beschrieben werden kann. Bei dieser handelt es sich um die zweimalige Abfolge von Photoionisation und Auger-Zerfall. Somit werden vier positive Ladungen im Molekül erzeugt. Das zweite Photon des XFEL wird dabei während der Dissoziation der sich Coulomb-abstoßenden Fragmente absorbiert, weshalb es sich um einen zweistufigen Prozess aus Anrege- und Abfrage- Schritt (Pump-Probe) handelt. Schlussendlich gelang zudem der Nachweis von Doppelinnerschalen-Vakanzen im Sauerstoff-Molekül nach Selektion des O2+/O2+- Aufbruchkanals. Hierfür konnten die beiden Möglichkeiten einer zweiseitigen oder einseitigen Doppelinnerschalen-Vakanz getrennt betrachtet werden und ebenfalls erstmalig das Verhalten der Elektronenemission dieser beiden Zustände verglichen werden.
Stellenwert der Teststreifen-basierten Analyse der INR für die Behandlung von Blutungskomplikationen

Das Ziel der hier vorliegenden Studie war es einen Zusammenhang zwischen den Ergebnissen von konventioneller versus Teststreifen-basierter INR-Messung zu untersuchen und die Analysedauern der beiden Methoden zu vergleichen. Wir haben in dieser prospektiven Mono-Center Studie 24 hämorrhagische Patienten und Patientinnen inkludiert und aus infrastrukturellen Gesichtspunkten in zwei Gruppenkollektive aufgeteilt. Das eine Studienkollektiv bildeten 12 hämorrhagische Patientinnen der Klinik für Gynäkologie und Geburtshilfe des Universitätsklinikums Frankfurt. Die Blutproben dieser Patientinnen wurden mittels einem personengebundenen Transportdienst in das Zentrallabor der Universitätsklinik geliefert. Das zweite Gruppenkollektiv bildeten 12 Patienten aus dem Schockraum der zentralen Notaufnahme. Die Blutproben dieses Kollektivs wurden mittels Rohrpost direkt in das Zentrallabor übermittelt. Wir untersuchten mittels konventioneller Gerinnungsdiagnostik und mittels Teststreifen-basierter POC-Diagnostik (CoaguChek II Pro®, PT Test, Roche Diagnostics AG) die INR eines jeden Patienten. Zudem erfolgte die Erfassung von Transport- und Analysedauer. Für die Auswertung der Daten errechneten wir die Spearman-Korrelationskoeffizienten sowohl auf Gruppenebene als auch für das Gesamtkollektiv und führten eine Bland-Altman Analyse zum direkten Methodenvergleich durch.
Es zeigte sich, dass die mittels POCT ermittelte INR im Gesamtkollektiv signifikant mit den im Zentrallabor gemessenen Werten korreliert (r=0,79). Auch auf Gruppenebene zeigte sich in Gruppe 1 (Schockraum) r=0,91 und in Gruppe 2 (Kreißsaal) r=0,83 eine signifikante Korrelation. Die Bland-Altmann Analyse ergab, dass die Ergebnisse der Teststreifen-basierten POC-Methode um 0,082 (SD±0,19) niedriger waren als die Ergebnisse der konventionellen Gerinnungstests. Die Untersuchung der Analyse- und Transportzeiten brachte hervor, dass die Bereitstellungsdauer der POC-Messmethode signifikant kürzer war (2 (1,04/2,85) Minuten) als die Dauer bis zur elektronischen Ergebnisbereitstellung nach laboranalytischen Untersuchungen (58,2 (38,28/88) Minuten). Es ergab sich zudem, dass die Transportdauer mittels Rohrpost mit 8 (3,25/10,1) Minuten signifikant kürzer war als die des personengebundenen Transportdienstes 18,5 (14,5/33) Minuten (p<0,001).Die in der Studie ermittelten konsistenten Ergebnisse lassen vermuten, dass Teststreifen-basierte Systeme als Methoden zur Notfalldiagnostik hämorrhagischer Patienten geeignet sein können, weil ihre Messergebnisse verglichen mit der klassischen Gerinnungsdiagnostik im Zentrallabor deutlich schneller und mit vergleichbarer Ergebnisqualität vorliegen. Die Teststreifen-basierten Methoden können als diagnostische Elemente in Hämotherapie-Algorithmen eingesetzt werden und dazu beitragen, eine zeitnahe und zielgerichtete Hämotherapie umzusetzen, die sich positiv auf das klinische Ergebnis der Patienten auswirken kann.
Das Gehirn weist in mehreren Bereichen anatomische Asymmetrien zwischen beiden Hemisphären auf, so auch in Bereichen der Hörrinde. Zudem ist bereits langjährig bekannt, dass menschliche Sprache vorrangig in der linken Gehirnhälfte, d.h. linksseitig lateralisiert, verarbeitet wird. Daraus folgend stellt sich die Frage, ob dies eine besondere Spezialisierung ist, oder ob es noch weitere lateralisierte Hirnfunktionen gibt. Viele akustische Signale haben dabei frequenzmodulierte (FM) Komponenten, die im Hörsystem für die Erkennung nach Parametern wie Richtung und Dauer der Modulation analysiert werden müssen. Ob die Analyse von FM-Komponenten oder einzelner Reizparameter im Gehirn lateralisiert stattfindet, wurde in der Literatur meist mit bildgebenden Verfahren untersucht.
Für das Erkennen und Unterscheiden der Modulationsrichtung weist eine Vielzahl von Studien auf eine erhöhte Aktivität in der rechten Hörrinde hin. Für die Analyse von Stimulusdauern ist es bisher allerdings noch unklar bzw. umstritten, ob diese lateralisiert erfolgt. Für die Untersuchung der Lateralisierung einfacher Sprachkomponenten werden häufig Konsonant-Vokal-Silben (CV-Silben) verwendet. In einer Vielzahl von Studien konnte eine linkslastige Lateralisierung, wie bei der Spracherkennung, gezeigt werden.
In der vorliegenden Arbeit wurde nun untersucht, ob ein eindeutigeres Muster von Lateralisierung zu finden ist, wenn diese in Wahrnehmungsexperimenten, untersucht wird. Dabei wurde ein zu untersuchender Teststimulus (FM-/CV-Stimulus) auf einem Ohr mit einem kontralateralen breitbandigen Rauschen auf dem anderen Ohr gleichzeitig präsentiert. Durch die Struktur der Hörbahn kann dabei davon ausgegangen werden, dass in einer Hemisphäre des Vorderhirns vorrangig Informationen aus dem kontralateralen Ohr verarbeitet und Informationen aus dem ipsilateralen Ohr unterdrückt werden und sich somit Rückschlüsse auf die Funktion/Beteiligung einer Hemishpäre ziehen lassen. Das Rauschen diente dabei zur unspezifischen Aktivierung der gegenüberliegenden Hemisphäre.
Die Lateralisierung wurde systematisch für unterschiedlich komplexe Reize untersucht. Dazu wurden in zwei Versuchsreihen Unterscheidungsexperimente durchgeführt, die sich in mehrere Messungen (mit mehreren Durchläufen) mit unterschiedlichen Parametereinstellungen gliederten. Pro Durchlauf musste sich die Versuchsperson immer zwischen zwei Antwortmöglichkeiten entscheiden (2-AFC-Verfahren). Der Schalldruckpegel des Rauschens war dabei für alle Messungen konstant. Der Schalldruckpegel der Teststimuli blieb zwar während einer Messung konstant, wurde jedoch innerhalb eines Experimentes von Messung zu Messung reduziert.
In einer gemeinsamen Analyse wurden jeweils die Fehlerraten und Reaktionszeiten beider Ohren, getrennt nach Seite und FM-/ CV-Stimulus, miteinander verglichen, um so auf eine mögliche Lateralisierung schließen zu können. Damit die Daten der Versuchspersonen bei vergleichbarer Schwierigkeit analysiert werden konnten, wurde als Vergleichswert zwischen allen Versuchspersonen der Schalldruckpegel der ersten Messung mit einer Fehlerrate von mindestens 15,0 % gewählt (15 %-Kriterium). Um auszuschließen, dass das Hörvermögen der Versuchspersonen Unterschiede zwischen beiden Ohren aufweist, wurde vor jeder Messung der „Punkt subjektiver Gleichheit“ für die Lautstärke-wahrnehmung zwischen linkem und rechten Ohr bestimmt.
In der ersten Versuchsreihe wurde dabei die Verarbeitung der Modulationsrichtung und der Stimulusdauer von FM-Stimuli untersucht. Es zeigte sich für beide Experimente, dass ein sinkender Schalldruckpegel des FM-Stimulus zu einer steigenden Fehlerrate führte. Unter Anwendung des 15 %-Kriteriums waren die Fehlerraten für die Unterscheidung der Modulationsrichtung signifikant geringer, wenn der FM-Stimulus auf dem linken Ohr präsentiert wurde. Dies ist ein deutlicher Hinweis für eine rechtslastige Lateralisierung.
Für die Unterscheidung der Stimulusdauer gab es dagegen keinen signifikanten Unterschied zwischen den Fehlerraten beider Ohren. Somit muss davon ausgegangen werden, dass beide Hemisphären für diese Aufgabe benötigt werden und eine bilaterale Verarbeitung stattfindet. In den Reaktionszeiten konnten in beiden Experimente keine signifikanten Unterschiede gezeigt werden. Die Unterscheidung der Modulationsrichtung wurde dabei von allen Versuchspersonen als einfacher eingestuft als die Unterscheidung der Stimulusdauer, was sich auch in niedrigeren Antwortschnelligkeit und Fehlerraten bei vergleichbaren Schalldruckpegeln zeigte.
In der zweiten Versuchsreihe wurde als Referenzmessung nochmals die Unterscheidung der Modulationsrichtungen von FM-Stimuli durchgeführt. Anschließend wurde die Unterscheidung von „da“ und „ga“ untersucht. Diese CV-Silben differieren ausschließlich in der FM-Komponente. Die Untercheidung von CV-Silben ohne Unterschied in der FM-Komponente wurde mittels „ta“ und „ka“ getestet. Für alle drei Experimente zeigte sich, dass ein geringerer Schalldruckpegel des FM- oder CV-Stimulus zu einer steigenden Fehlerrate führte. Unter Anwendung des 15 %-Kriteriums zeigte sich für die Unterscheidung der Modulationsrichtung ein Trend zu niedrigeren Fehlerraten bei der Präsentation des FM-Stimulus auf dem linken im Vergleich mit dem rechten Ohr. In den Reaktionszeiten konnten keine signifikanten Unterschiede gezeigt werden.
Für die Unterscheidung von „da“ und „ga“ ließ sich unter Anwendung des 15 %-Kriteriums in den Fehlerraten und Reaktionszeiten kein Vorteil eines Ohres nachweisen. Dagegen zeigten sich klare Unterschiede bei einzelnen Versuchspersonen. So waren die Fehlerraten für Versuchspersonen, die vorwiegend „da“ erkannt bzw. gehört hatten signifikant höher, wenn der CV-Stimulus auf dem rechten Ohr präsentiert wurde, für „ga“-Hörer war das Gegenteil der Fall. In den Reaktionszeiten konnte kein signifikanter Zusammenhang nachgewiesen werden. Somit ließ sich zeigen, dass je nach Strategie der Versuchsperson bzw. deren individueller Wahrnehmung der CV-Silben, Unterschiede in der Lateralisierung erreicht werden können.
Für die Unterscheidung von „ta“ und „ka“ zeigten sich unter Anwendung des 15 %-Kriteriums signifikant niedrigere Fehlerraten und Reaktionszeiten, wenn der CV-Stimulus auf dem linken Ohr präsentiert wurde. Dies weist deutlich auf eine rechtslastige Lateralisierung hin. Vergleicht man alle drei Experimente ließ sich zudem zeigen, dass die Unterscheidung der Modulationsrichtung einfacher war als die Unterscheidung verschiedener CV-Stimuli. Dabei war die Unterscheidung von „da“ und „ga“ für die Versuchspersonen schwieriger als die Unterscheidung von „ta“ und „ka“. Allerdings konnte in den Lateralisierungsdaten kein direkter Zusammenhang zwischen den FM- und „da“-/„ga“-Stimuli gezeigt werden.
Zusammenfassend konnte in allen fünf Experimenten eine verschieden stark lateralisierte Verarbeitung von akustischen Stimuli bei gleichzeitigem kontralateralen Rauschen gezeigt werden. Der Vorteil eines Ohres (bzw. einer Hemisphäre) war sowohl von der Aufgabe als auch vom Stimulustyp abhängig. Dabei gab es zum Teil starke Unterschiede in der Effektstärke und dem Grad der Lateralisierung zwischen den einzelnen Versuchspersonen. Insgesamt konnte gezeigt werden, dass sich die hier angewendete psychophysische Methode gut eignet, um Ergebnisse zur Lateralisierung von akustischen Stimuli zu gewinnen und somit die Verhaltensrelevanz von Ergebnissen aus Studien mit bildgebenden Verfahren zu überprüfen.

The application of natural products (NPs) as drugs and lead compounds has greatly improved human health over the past few decades. Despite their success, we still need to find new NPs that can be used as drugs to combat increasing drug resistance via new modes of action and to develop safer treatments with less side effects.
Entomopathogenic bacteria of Xenorhabdus and Photorhabdus that live in mutualistic symbiosis with nematodes are considered as promising producers of NPs, since more than 6.5% of their genomes are assigned to biosynthetic gene clusters (BGCs) responsible for production of secondary metabolites. The investigation on NPs from Xenorhabdus and Photorhabdus can not only provide new compounds for drug discovery but also help to understand the biochemical basis involved in mutualistic and pathogenic symbiosis of bacteria, nematode host and insect prey.
Nonribosomal peptides (NRPs) are a large class of NPs that are mainly found in bacteria and fungi. They are biosynthesized by nonribosomal peptide synthetases (NRPSs) and display diverse functions, representing more than 20 clinically used drugs. Although a large number of NRPs have been identified in Xenorhabdus and Photorhabdus, the advanced genome sequencing and bioinformatic analysis indicate that these bacteria still have many unknown NRPS-encoding gene clusters for NRP production that are worth to explore. Therefore, this thesis focuses on the discovery, biosynthesis, structure identification, and biological functions of new NRPs from Xenorhabdus and Photorhabdus.
The first publication describes the isolation and structure elucidation of seven new rhabdopeptide/xenortide-like peptides (RXPs) from X. innexi, incorporating putrescine or ammonia as the C-terminal amines. Bioactivity testing of these RXPs revealed potent antiprotozoal activity against the causative agents of sleeping sickness (Trypanosoma brucei rhodesiense) and malaria (Plasmodium falciparum), making them the most active RXP derivatives known to date. Biosynthetically, the initial NRPS module InxA might act iteratively with a flexible methyltransferase activity to catalyze the incorporation of the first five or six N-methylvaline/valine to these peptides.
The second publication focuses on the structure elucidation of seven unusual methionine-containing RXPs that were found as minor products in E. coli carrying the BGC kj12ABC from Xenorhabdus KJ12.1. To confirm the proposed structures from detailed HPLC-MS analysis, a solid-phase peptide synthesis (SPPS) method was developed for the synthesis of these partially methylated RXPs. These RXPs also exhibited good effects against T. brucei rhodesiense and P. falciparum, suggesting RXPs might play a role in protecting insect cadaver from soil-living protozoa to support the symbiosis with nematodes.
The third publication presents the identification of a new peptide library, named photohexapeptide library, which occurred after the biosynthetic gene phpS was activated in P. asymbiotica PB68.1 via promoter exchange. The chemical diversity of the photohexapeptides results from unusual promiscuous specificity of five out of six adenylation (A) domains being an excellent example of how to create compound libraries in nature. Furthermore, photohexapeptides enrich the family of the rare linear D-/L-peptide NPs.
The fourth publication concentrates on the structure elucidation of a new cyclohexapeptide, termed photoditritide, which was produced by P. temperata Meg1 after the biosynthetic gene pdtS was activated via promoter exchange. Photoditritide so far is the only example of a peptide from entomopathogenic bacteria that contains the uncommon amino acid homoarginine. The potent antimicrobial activity of photoditritide against Micrococcus luteus implies that photoditritide can protect the insect cadaver from food competitor bacteria in the complex life cycle of nematode and bacteria.
The last publication reports a new family of cyclic lipopeptides (CLPs), named phototemtides, which were obtained after the BGC pttABC from P. temperata Meg1 was heterologously expressed in E. coli. The gene pttA encodes an MbtH protein that was required for the biosynthesis of phototemtides in E. coli. To determine the absolute configurations of the hydroxy fatty acids, a total synthesis of the major compound phototemtide A was performed. Although the antimalarial activity of phototemtide A is only weak, it might be a starting point towards a selective P. falciparum compound, as it shows no activity against any other tested organisms.
