The ribonucleic acid of reovirus was extracted with 2 M sodium perchlorate solution and spread by the protein monolayer technique. Areas of the monolayer were transferred to support films, rotary shadowed, and observed in the electron microscope. Filaments of RNA obtained by extraction prior to spreading were similar in appearance and in distribution of contour lengths (0.2 to 1.2 μ) to those obtained by phenol extraction of the virus. Most of the filaments resulting from extraction of the virus suspension during spreading on a sodium perchlorate solution, however, were longer than 1 μ. The lengths of the longest filaments exceeded the 5 μ length predicted from chemical data for one single piece of complementary-stranded RNA in the reovirus particle.
The short filaments, 1.2 μ and less in length, fell into a tri-modal pattern of length distribution with peaks at 0.35 μ, 0.60 μ and 1.10 μ. These shorter lengths probably resulted from breakage of the intact RNA during the extraction procedures. The consistently observed pattern of length distribution suggests that they represent relatively stable subunits of the molecule.
Sodium perchlorate extracted reovirus RNA was thermally denatured in formaldehyde prior to spreading by the protein monolayer technique. Length distributions and relative numbers of filaments in the peaks of the tri-modal distribution pattern were similar to those found for unheated material when extracted prior to spreading. This similarity indicates that heating subsequent to extraction produced no further filament breakage. The thin, kinky appearance of the heated filaments, and the appearance of congruent pairs, indicated that heating had separated the strands of the complementary-stranded RNA subunits.
The title compound, C26H18BrNO4, features a functionalized chromene. The cyclohexene ring adopts a sofa conformation and has the nitro group and the bromophenyl ring in an axial position. The ten atoms of the chromene moiety lie close to a common plane (r.m.s. deviation = 0.066 Å). The attached phenyl ring is twisted by 32.89 (10)° from the chromene plane. The crystal packing is stabilized by C—H[cdots, three dots, centered]O interactions.
3,17 β-Hydroxysteroid dehydrogenase has been enriched and purified from cytosol of Streptomyces hydrogenans. After ammonium sulfate precipitation and filtration on Sephadex G-100 the enzyme was finally purified by preparative gel electrophoresis and DEAE-Sephadex A-50 chromatography. Polyacrylamide gel electrophoresis in the presence of sodium dodecylsulfate gave a single band of mobility corresponding to molecular weight of 70 200 ± 2 500. 3 β-. 17 β- as well as 20 β-hydroxy steroids were dehydrogenated by the enzyme in the presence of NAD+. The dehydrogenation proceeded faster than the reduction of the corresponding ketosteroids in the presence of NADH. The enzyme does not accent NADP+ or NADPH as co-substrates. The apparent Km values were calculated to be 11 μᴍ for 5 α-dihydrotestosterone, 20 μᴍ for testosterone ana 68 μᴍ for epiandrosterone in the NAD+-driven reaction, 1.8 x 10-4 m for NADH+ and 1.9 x 10-4 ᴍ for NADH. The catalytic activity was influenced by the ratio of NAD+/ATP. The inhibition by ATP appears to be of a competitive type with respect to NAD+ (Ki 1.15 x 10-3 ᴍ).
After sucrose gradient centrifugation in a preparative ultracentrifuge the enzyme sediments with 4.1 ± 0.1 S as estimated in comparison to other proteins of known sedimentation coefficient. The isoelectric point was determined to be 3.9 with the LKB preparative isoelectric focusing column (pH 2-11) and 4.1 with the analytical flat bed polyacrylamide isofocusing (pH 3 - 5). The number of SH groups was determined to be 2 mol/mol enzyme. In the presence of 6 M urea the figure inceases to 3 mol SH/mol enzyme. In the presence of an excess of p-chloromercuribenzoate the enzyme activity decreases only partially.
Although in general cells are genetically identical in multicellular organisms, the differential expression of genomic information enables cell type definition and specific organ function. In eukaryotic cells, the DNA is associated with histone and non-histones proteins into a restrictive structure called chromatin. Assembly into chromatin does not only protect and package the linear double stranded DNA into the nucleus but is fundamental for the execution of diverse genetic programs. Posttranslational modifications of histones regulate the accessibility of the DNA to transcription factors and serve as scaffold for binding of regulatory proteins. Nuclear receptors are transcription factors that bind specific target sequences on the DNA and recruit transcriptional coregulators at the promoter. These are able to modify the chromatin structure in an activating or repressing manner. The contribution of corepressors to the biological actions of nuclear receptors has turned out to be essential. Impaired corepressor function can be the cause of endocrine malfunctions, neoplastic diseases or severe developmental abnormalities. To better understand the role of the nuclear receptor corepressor N-CoR the unknown function of the extreme C-terminus was investigated. In this thesis the interaction of N-CoR with the non-POU-domain containing octamer-binding protein Non0/p54nrb, that was found tobe a potential interaction partner in a yeast-two-hybrid screen, was confirmed. This protein contains two RNA recognition motifs (RRM) and is described as a multifunctional protein since it is involved in transcription Initiation as well as in pre-mRNA processing. The RRM1 motif was determined to be essential and sufficient for the interaction with N-CoR. Obtaining dominant negative effect with the Non0/p54nrb RRM1 deletion mutant in functional reporter assays, data support that NonO modulates the capacity of N-CoR to repress and alters the recruitment of N-CoR by nuclear receptors to targeted Promoters. Additional analyses suggest that the N- and C- terminus of N-CoR are involved in intramolecular interactions and that they regulate each other. Taken results together a functional model is proposed that supports the biological relevance of the interaction of N-CoR with NonO and the function of N-CoR C-terminus acting as asensor that evaluates the ratio of corepressors and coactivators in the nuclear receptor environment. N-CoR repressive capacity would be altered by modulating factors like NonO that interacts with N-CoR C-terminus. The mechanism support that splicing and transcription regulation are physically and functionallylinked to ensure the appropriate amount of messager RNA to be transcript and process in response to stimulation intensity and cell context.
Mitochondrial complex I, the largest and most complicated proton pump of the respiratory chain, links the electron transfer from NADH to ubiquinone to the pumping of four protons from the matrix into the intermembrane space. In humans, defects in complex I are involved in a wide range of degenerative disorders. Recent progress in the X-ray structural analysis of prokaryotic and eukaryotic complex I confirmed that the redox reactions are confined entirely to the hydrophilic peripheral arm of the L-shaped molecule and take place at a remarkable distance from the membrane domain. While this clearly implies that the proton pumping within the membrane arm of complex I is driven indirectly via long-range conformational coupling, the molecular mechanism and the number, identity, and localization of the pump-sites remains unclear. Here, we report that upon deletion of the gene for a small accessory subunit of the Yarrowia complex I, a stable subcomplex (nb8m delta) is formed that lacks the distal part of the membrane domain as revealed by single particle analysis. The analysis of the subunit composition of holo and subcomplex by three complementary proteomic approaches revealed that two (ND4 and ND5) of the three subunits with homology to bacterial Mrp-type Na+/H+ antiporters that have been discussed as prime candidates for harbouring the proton pumps were missing in nb8m delta. Nevertheless, nb8m delta still pumps protons at half the stoichiometry of the complete enzyme. Our results provide evidence that the membrane arm of complex I harbours two functionally distinct pump modules that are connected in series by the long helical transmission element recently identified by X-ray structural analysis.
Air pollution of particulate matter (PM) from traffic emissions has a significant impact on human health. Risk assessments for different traffic participants are often performed on the basis of data from local air quality monitoring stations. Numerous studies demonstrated the limitation of this approach. To assess the risk of PM exposure to a car driver more realistically, we measure the exposure to PM in a car cabin with a mobile aerosol spectrometer in Frankfurt am Main under different settings (local variations, opened versus a closed window) and compare it with data from stationary measurement. A video camera monitored the surroundings for potential PM source detection. In-cabin concentrations peaked at 508 µg m−3 for PM10, 133.9 µg m−3 for PM2.5, and 401.3 µg m−3 for coarse particles, and strongly depended on PM size and PM concentration in ambient air. The concentration of smaller particles showed low fluctuations, but the concentration of coarse particles showed high fluctuations with maximum values on busy roads. Several of these concentration peaks were assigned to the corresponding sources with characteristic particle size distribution profiles. The closure of the car window reduced the exposure to PM, and in particular to coarse particles. The mobile measured PM values differed significantly from stationary PM measures, although good correlations were computed for finer particles. Mobile rather than stationary measurements are essential to assess the risk of PM exposure for car passengers.
Sauerstoff wird in vielen prokaryotischen und eukaryotischen Atmungsketten von der Cytochrom c Oxidase zu Wasser reduziert. Dieses Enzym besteht in Paracoccus denitrificans aus vier Untereinheiten, von denen aber nur die ersten zwei für die Funktion essentiell sind. Elektronen werden von einem membrangebundenen Cytochrom c552 zunächst auf das CuA-Zentrum der Untereinheit II der Oxidase übertragen und gelangen von dort zum low-spin Häm a der Untereinheit I. Am binukleären Zentrum, das aus dem high-spin Häm a3 und dem CuB-Atom besteht, erfolgt schließlich die Reduktion von molekularem Sauerstoff zu Wasser. Hauptziel dieser Arbeit war es, die Bindungsstelle für Cytochrom c durch das gezielte Einführen oder Entfernen negativer Ladungen auf der Oxidase-Oberfläche zu bestimmen. Die Charakterisierung dieser Mutanten erfolgte hauptsächlich durch spektroskopische Analyse der kinetischen Parameter unter Umsatzbedingungen. Zusätzlich wurde der Teilschritt der Susbtratbindung durch die Aufnahme von Assoziationskinetiken untersucht. Als Substrat diente das reduzierte mitochondriale Cytochrom c oder ein lösliches Fragment des Paracoccus-Cytochrom c552. Die eingeführten Mutationen beeinflußten in unterschiedlicher Weise die Bindung der beiden verwendeten Substrate. Hierbei ist der negativ geladene Bereich der Oberfläche von Untereinheit II der Oxidase, der zur Bindung des Paracoccus-eigenen Substrats beiträgt, größer als der für die Wechselwirkung mit dem mitochondrialen Cytochrom c. Erstmals kann somit ein Modell für die Bindung des nativen Substrats an die Paracoccus-Oxidase aufgestellt werden. Die aa3-Oxidase aus Paracoccus zeigte keine Elektronentransfereaktion mit dem Cytochrom c552 aus Thermus thermophilus, da dessen Oberfläche nicht die hierfür erforderlichen Ladungen trägt. Durch Erhöhung der Hydrophobizität auf der Untereinheit II der Paracoccus-Oxidase wurde deren Oberfläche so verändert, dass sie nunmehr auch durch dieses „hydrophobe“ Cytochrom c reduziert werden konnte. Der Aminosäurerest W121 auf der Oberfläche der Untereinheit II wurde in der Vergangenheit als ein möglicher Elektroneneintrittsort in die Cytochrom c Oxidase beschrieben. Die in der vorliegenden Arbeit an dieser Position hergestellten Mutanten zeigten zwar drastische Einbußen in den maximalen Wechselzahlen, aber eine weitgehend unveränderte Affinität zum Cytochrom c. Die exakte Positionierung der Seitengruppe des Tryptophans spielt hierbei gegenüber der Aromatizität die entscheidende Rolle. An benachbarten Positionen eingeführte Mutationen ergaben keine Hinweise auf alternative Elektroneneintrittsorte. Durch ortsspezifische Mutagenese auf dem löslichen Cytochrom c552-Fragment wurden die Lysine auf der Oberfläche entfernt, um zu klären, ob die Wechselwirkung zwischen Cytochrom c und Oxidase auf Ladungspaaren beruht oder über das Gesamtpotential der Bindungsstellen bestimmt wird. Die Lysin-Mutanten zeigten alle im gleichen Maße eine reduzierte Affinität zur Oxidase. Die vorliegenden Ergebnisse lassen den Schluß zu, dass die Wechselwirkung zwischen Oxidase und Cytochrom c552 über das Gesamt-Oberflächenpotential vermittelt wird. Steady-state-Kinetiken unter Verwendung von Pferdeherz-Cytochrom c oder Paracoccus Cytochrom c552 weichen von den klassichen Michaelis-Menten-Verhalten ab, da zwei separate Phasen für die Kinetik beobachtet werden. Dieses Phänomen bisher wurde mit der Anwesenheit einer zweiten Bindungsstelle oder einer redoxgekoppelten Konformationsänderung der Oxidase erklärt. Durch Untersuchungen von Oxidase-Präparationen mit unterschiedlicher Untereinheiten-Zusammensetzung oder gebundenem Antikörper konnte gezeigt werden, dass die Biphasizität der Oxidase-Kinetik ! nicht auf der Anwesenheit der Untereinheiten III und IV beruht, ! durch die spezifische Bindung von Antikörpern zu niedrigeren Ionenstärken verschoben wird bei gleichzeitiger Verringerung der Affinität zum Substrat, ! eine intrinsische Eigenschaft aller untersuchten Mutanten ist. Im Rahmen dieser Untersuchungen konnten keine Hinweise auf eine zweite Cytochrom c Bindungsstelle gefunden werden. Ein weiteres Ziel dieser Arbeit war die Expression, Reinigung und Charakterisierung des homologe, membrangebundenen Cytochrom c552 aus P. denitrificans. Das Protein wurde mit einem Histidin-Tag versehen und heterolog in E. coli in guter Ausbeute exprimiert und gereinigt. Seine Charakterisierung über steady-state-Messungen ergab ein deutlich verschobenes pH-Optimum mit höherer Affinität zur Oxidase im Vergleich zu seinen verkürzten löslichen Fragmenten.
Regulation des matrizellulären Proteins SMOC-1 durch Zytokine und Stickoxid in Rattenmesangiumzellen

Zytokine stimulieren in Mesangiumzellen die Produktion und die Freisetzung großer Mengen entzündlicher Mediatoren. In dieser Arbeit wurden mit Hilfe der RAP-PCR („RNA arbitrarily primed polymerase chain reaction“), einer auf mRNA basierenden „Differential display“-Methode, die Effekte von Interleukin-1β (IL-1β) auf das Genexpressionsmuster in glomerulären Rattenmesangiumzellen untersucht. Dabei wurde das matrizelluläre Glykoprotein „Secreted modular calcium-binding protein-1“ (SMOC-1) identifiziert, welches in Mesangiumzellen durch IL-1β herunterreguliert wird. SMOC-1 wird von verschiedenen Zelltypen exprimiert und sezerniert, doch seine biologische Funktion konnte bisher nicht aufgedeckt werden. Weitere Experimente bestätigten, dass die mRNA- und Proteinexpression von SMOC-1 durch proinflammatorische Zytokine, wie IL-1β und Tumornekrosefaktor-α (TNF-α) herunterreguliert wird. Dieser Effekt wird zu einem großen Teil durch die endogene Freisetzung von Stickoxid (NO) aufgrund der Aktivität der induzierbaren NO-Synthase (iNOS) und der nachfolgenden Aktivierung der löslichen Guanylatzyklase (sGC) vermittelt. Außerdem tragen auch reaktive Sauerstoffver-bindungen (ROS), deren Bildung durch die Zytokine verstärkt wird, zur Herunter-regulierung der SMOC-1-Expression bei. Durch In-situ-Hybridisierungsexperimente konnte ferner gezeigt werden, dass die Hemmung der NO-Synthese durch den spezifischen iNOS-Inhibitor L-NIL in einem Rattenmodell der anti-Thy1.1-Glomerulonephritis die SMOC-1-Expression deutlich erhöhte. Dies belegt somit auch in vivo die biologische Relevanz von NO in der Modulierung der SMOC-1-Expression. Die funktionelle Rolle von SMOC-1 in Mesangiumzellen wurde durch die Hemmung der SMOC-1-Expression mit Hilfe einer spezifischen siRNA untersucht. Dabei zeigte sich, dass die Hemmung von SMOC-1 eine deutliche Inhibierung der mRNA-Expression von „Transforming growth factor β1“ (TGF-β1) sowie dessen Gesamtproteinspiegel zur Folge hat. Auch die Aktivität von TGF-β1 wurde reduziert, wie anhand der verringerten Spiegel an aktivem TGF-β1-Protein und der verringerten mRNA-Expression bekannter TGF-β-regulierter Gene, wie „Connective tissue growth factor“ (CTGF), „Plasminogen activator inhibitor-1“ (PAI-1) und Biglykan gezeigt wurde. Diese Ergebnisse deuten auf eine Rolle von SMOC-1 bei der Modulierung des TGF-β1-Signalwegs hin. Zusammenfassend betrachtet scheint NO die SMOC-1-Expression in der akuten glomerulären Entzündung zu vermindern und dadurch die TGF-β-getriebenen profibrotischen Signalprozesse zu limitieren. Der zweite Aspekt dieser Arbeit befasst sich mit der Rolle von Peroxisomen-Proliferator-aktivierten Rezeptoren (PPAR) in der IL-1β-vermittelten Expression der induzierbaren NO-Synthase. PPARα-Aktivatoren steigern in Mesangiumzellen die IL-1β-induzierte Aktivität der iNOS, während die Hemmung von PPARα durch spezifische Inhibitoren oder siRNA die iNOS-Expression/-Aktivität deutlich reduziert. Die Ergebnisse von Promotor-studien zeigten die essentielle Rolle einer möglichen PPAR-Bindestelle im iNOS-Promotor. IL-1β scheint die Bildung eines endogenen PPAR-Liganden zu induzieren, wodurch die Bindung von PPAR-Proteinkomplexen an das regulatorische DNA-Element im iNOS-Promotor verstärkt und dessen Aktivität gesteigert wird. Daneben scheinen jedoch auch Nebeneffekte der PPAR-Aktivatoren, wie die Freisetzung von ROS, zur synergistischen Wirkung auf die IL-1β-induzierte iNOS-Expression beizutragen. Die Wirkung von dualen PPARα/γ-Aktivatoren auf entzündliche Prozesse wird seit längerem diskutiert, daher wurden die biologischen Effekte von neu synthetisierten möglichen dualen PPARα/γ-Aktivatoren auf Entzündungsparameter, wie z. B. die iNOS-Expression, in Mesangiumzellen untersucht. Alle untersuchten Aktivatoren steigerten in der Form von Esterverbindungen die IL-1β-induzierte Expression der iNOS sowie der sekretorischen Phospholipase A2 (sPLA2), während die entsprechenden freien Säuren wenig Effekte zeigten. Diese proinflammatorische Wirkung scheint jedoch weniger auf einer Aktivierung des PPAR-Rezeptors zu beruhen als auf der Freisetzung von ROS, die durch die Aktivatoren teilweise deutlich erhöht wurde. Weitere Experimente zur Charakterisierung der PPAR-spezifischen Wirkung der Aktivatoren sowie zur optimalen Wirkkonzentration sind nötig, bevor der Effekt dieser Aktivatoren auf die inflammatorische Genexpression genau bewertet werden kann.
Hemoproteinoids related to contemporary porphyrin-dependent peroxidases were synthesized under simple conditions. The peroxidative activity of hematin increased by a factor of 50 if the hematin was bound to proteinoids whereas the catalatic activity of hematin decreased rather under the same conditions. The peroxidative activity of hemoproteinoids particularly increased with their lysine content whereas the catalatic activity especially decreased in proteinoids with high phenylalanine content. The isoelectric points of the lysine-rich peroxidic hemoproteinoids were about 8. Their relatively broad pH-activity optimum was about pH 7.0. The molecular weights were a little below 20 000. Hematin content and amino acid composition of the synthetic materials were varied greatly. The substrate specificity appeared as broad as that of biogenous peroxidases, e. g., horseradish peroxidase. Among the many substrates was NADH. The possible importance of the peroxidative oxidation of NADH-type coenzymes by primitive heterotrophic organisms or prebiological systems in an anaerobic environment is discussed.
The formation of aliphatic α-amino acids by X-ray induced carboxylation of simple amines or amination of simple carboxylic acids is not favored over the formation of other amino acids. The new carboxylic and amino groups are more or less distributed statistically over the carbon atoms of the starting material. On radiationchemical formation of aliphatic hydrocarbons over C3, therefore, an increasing amount of unusual amino acids is produced. The results are influenced by various parameters such as temperature, pH, concentration, linear energy transfer and total dosis of radiation applied. Also peptides can be formed radiationchemically. However, the formation of greater molecules by radiationchemical processes under the conditions of a primitive earth seems to have only a low probability. The reaction mechanisms of radiationchemical carboxylation and amination are discussed.
