The title compound, [Fe2(C5H5)2(C24H22BP2)(CO)4][FeCl4]·CHCl3, is an oxidation product of CpFe(CO)2PPh2BH3. One pair of phenyl rings attached to the two different P atoms are almost parallel, as are the other pair [dihedral angles = 8.7 (5) and 8.9 (5)°]. The planes of the two cyclopentadienyl rings are inclined by 26.8 (7)° with respect to each other. The carbonyl groups at each Fe atom are almost perpendicular [C-Fe-C = 92.6 (6) and 94.3 (5)°]. Key indicators: single-crystal X-ray study; T = 173 K; mean σ(C–C) = 0.019 Å; R factor = 0.112; wR factor = 0.177; data-to-parameter ratio = 16.8.
The geometric parameters of the molecule of the title compound, C14H16O2P2, are in the usual ranges. It is a meso compound with the two chiral P atoms having opposite configurations. The P-CH2-CH2-P chain adopts a trans conformation [torsion angle -178.59 (17)°]. The P=O bonds are almost coplanar with the adjacent phenyl ring [torsion angles = 3.8 (3) and 0.3 (3)°]. Whereas one of them is synclinal [torsion angle = -59.0 (2)°] to the central C-C bond, the other is anticlinal [torsion angle = 56.6 (2)°] to the central C-C bond. The dihedral angle between the two phenyl rings is 5.2 (3)°. The molecules are linked by weak C-H...O hydrogen bonds. They crystallize in rows running along the c axis. Key indicators: single-crystal X-ray study; T = 173 K; mean σ(C–C) = 0.005 Å; R factor = 0.038; wR factor = 0.093; data-to-parameter ratio = 15.2.
Decades of work have demonstrated that mRNAs are localized and translated within neuronal dendrites and axons to provide proteins for remodeling and maintaining growth cones or synapses. It remains unknown, however, whether specific forms of plasticity differentially regulate the dynamics and translation of individual mRNA species. To address these issues, we targeted three individual synaptically-localized mRNAs, CamkIIa, Beta actin, Psd95, and used molecular beacons to track endogenous mRNA movements and reporters and Crispr-Cas9 gene editing to track their translation. We found widespread alterations in mRNA behavior during two forms of synaptic plasticity, long-term potentiation (LTP) and depression (LTD). Changes in mRNA dynamics following plasticity resulted in an enrichment of mRNA in the vicinity of dendritic spines. Both the reporters and tagging of endogenous proteins revealed the transcript-specific stimulation of protein synthesis following LTP or LTD. The plasticity-induced enrichment of mRNA near synapses could be uncoupled from its translational status. The enrichment of mRNA in the proximity of spines allows for localized signaling pathways to decode plasticity milieus and stimulate a specific translational profile, resulting in a customized remodeling of the synaptic proteome.
Decades of work have demonstrated that messenger RNAs (mRNAs) are localized and translated within neuronal dendrites and axons to provide proteins for remodeling and maintaining growth cones or synapses. It remains unknown, however, whether specific forms of plasticity differentially regulate the dynamics and translation of individual mRNA species. Studies over the past decade have revealed that metabolism profoundly influences immune responses. In particular, metabolism causes epigenetic regulation of gene expression, as a growing number of metabolic intermediates are substrates for histone post-translational modifications altering chromatin structure. One of these substrates is acetyl-coenzyme A (CoA), which donates an acetyl group for histone acetylation. Cytosolic acetyl-CoA is also a critical substrate for de novo synthesis of fatty acids and sterols necessary for rapid cellular growth. One of the main enzymes catalyzing cytosolic acetyl-CoA formation is ATP-citrate lyase (ACLY). In addition to its classical function in the provision of acetyl-CoA for de novo lipogenesis, ACLY contributes to epigenetic regulation through histone acetylation, which is increasingly appreciated. In this review we explore the current knowledge of ACLY and acetyl-CoA in mediating innate and adaptive immune responses. We focus on the role of ACLY in supporting de novo lipogenesis in immune cells as well as on its impact on epigenetic alterations. Moreover, we summarize alternative sources of acetyl-CoA and their contribution to metabolic and epigenetic regulation in cells of the immune system.
In der Arbeit wurden neue Komplexbildner für die nasschemische, alkalische Reinigung von Siliciumhalbleiteroberflächen vorgestellt. Dabei handelte es sich neben kommerziell verfügbaren Verbindungen, wie beispielsweise Tiron, um den Chelatbildner Pyrinan und die vielversprechende Gruppe der 3-Hydroxypyridin-4(1H)-on-Komplexbildner („Pyridinone“). Insbesondere Letztere sind als effiziente Eisenkomplexbildner von präklinischen Untersuchungen zur Therapie von Eisenstoffwechselerkrankungen bekannt und haben deshalb großes Interesse hervorgerufen. Die Darstellung von Pyrinan und der Pyridinone war in befriedigenden bis guten Ausbeuten mit herkömmlichen Syntheseschritten möglich. Die Synthese konnte somit die im Abschnitt 1.2.1 erwähnten Anforderungen an die Komplexbildner befriedigen. Hinsichtlich der an die Strukturierung von Halbleitermaterialen geknüpften, jedoch bei der Präparation der Komplexbildner nicht realisierten Reinheitsbedingungen, wiesen diese einen teilweise erheblichen Kontaminationsgrad durch Metalle auf. Demzufolge hätten die Komplexbildner als potentielle Kontaminationsquellen bei der SC1-Reinigung von Siliciumsubstraten wirken können. Letzteres konnte jedoch im Rahmen der zehnminütigen Immersionsdauer in ¼/20 SC1 bei 70 °C weitgehend ausgeschlossen werden. Der Trend bei der nasschemischen Reinigung von Halbleitersubstraten mit immer stärker verdünnten Reinigunslösungen und reduzierten Reinigungsbadtemperaturen zu arbeiten, sollten dieses und die folgend zusammengefassten Ergebnisse noch weiter begünstigen. Es wurden Reinigungsexperimente mit Siliciumsubstraten und mit den Reinigungslösungen absichtlich zugesetzten Kontaminanten durchgeführt, um die Wirkungen der einzelnen Verbindungen besser studieren zu können. Es wurden die Metalle Al, Fe, Zn, Cu, Ni und Cr, die in der Halbleiterproduktion als gängige Kontaminanten auftreten, untersucht. Die Metalle Zn, Ni und Cu spielen in der SC1-Lösung, mit abnehmender Gewichtung in der genannten Reihenfolge, aufgrund deren Amminkomplexbildung keine nennenswerten Rolle als Oberflächenkontaminanten. Cr(III) wird in der SC1-Lösung zu Chromat oxidiert und kontaminiert aus diesem Grund die Oberfläche ebenfalls nicht. Dem gegenüber sind Al und Fe unter den gewählten Bedingungen sehr starke Oberflächenkontaminanten. Prinzipiell eignen sich fast alle untersuchten Komplexbildner, die Siliciumoberfläche vor der Kontamination durch Fe aus SC1 zu schützen. Hinsichtlich ihrer Wirksamkeit gegenüber Al und Zn wurden sie jedoch deutlich diskriminiert. Daraus schlussfolgernd ist der Einsatz mehrerer Komplexbildnern erforderlich, um die Kontaminationsgefahr der Siliciumoberfläche durch alle genannten Metalle gezielt kontrollieren und über einen gewissen Schwellenwert vermeiden zu können. Die Wirkung von Pyrinan und der Pyridinone ist vergleichbar, meistens jedoch deutlich besser, als die herkömmlicher Poly(alpha-aminomethylencarbonsäuren) (z. B. EDTA, Untersuchungen von IMEC). Sie erzielen Oberflächenkonzentrationen kritischer Kontaminanten (z. B. Fe) in der Größenordnung, die für die Prozessierung aktueller und künftiger Generationen von Bauelementen erforderlich sind. Konsequenterweise sind deshalb sowohl Pyrinan als auch die Pyridinonkomplexbildner und deren Verwendungszweck zum Patent angemeldet worden. In einem weiteren Teil der Arbeit wurde die Eignung der Komplexbildner bestimmt, hochkonzentrierte, den Halbleiterspezifikationen entsprechende Lösungen von H2O2 stabilisieren zu können. Die Stabilität wurde in Abhängigkeit von drei vorgegebenen Lagerungstemperaturen verfolgt. Sowohl die vorgestellten Verbindungen als auch die als Referenzsubstanz mituntersuchte Verbindung Dequest® 2060s konnten die an einen geeigneten Stabilisator gestellten Bedingungen nicht befriedigen. Lediglich die nicht stabilisierte Lösung von H2O2, die Referenzprobe, besaß eine, über den gesamten Beobachtungszeitraum ausreichende Stabilität. Das mangelhafte Abschneiden der vorgestellten Verbindungen als potentielle Stabilisatoren wird deren partieller bzw. vollständiger Oxidation durch H2O2 zugeordnet und ist am Beispiel von Dequest® 2060s weiter diskutiert worden. Im Zuge der Oxidation werden die intrinsischen Metallkontaminationen der Komplexbildner freigesetzt, wodurch die Kontaminanten in die Lage versetzt werden können, in das Reaktionsgeschehen katalytisch einzugreifen und die Zersetzung der Proben zu beschleunigen. Die Lagerung der Komplexbildner in konzentriertem NH3 stellt die Alternative zur Lagerung in H2O2 dar, wenn man zu einem fertig einsetzbaren, industriellen Produkt für die nasschemische Reinigung von Halbleiteroberflächen gelangen möchte, welches eine einstufige alkalische Reinigung (APM+®) ermöglicht. Hier konnten bis auf die Proben, welche Tiron als Komplexbildner enthielten, keine Besonderheiten festgestellt werden. Die mit Tiron versetzten Proben führen bereits nach sehr kurzer Lagerungsdauer zu einer intensiv rot gefärbten Lösung, die der im Alkalischen und durch Luftsauerstoff erfolgenden Oxidation von Tiron durch Luftsauerstoff zugerechnet wird. Eine, die Reinigungsexperimente ergänzende und wichtige Information, stellt die Stabilität der Komplexbildner und durch diese bedingt die der gesamten Reinigungslösung dar. Letztere wurde durch die titrimetrische Bestimmung der Konzentration von H2O2 ausgesuchter Reinigungslösungen bestimmt. Die Stabilität des Komplexbildners DEHP wurde stellvertretend für die ganze Gruppe (der einfachen Pyridinone) in verschiedenen, in der Halbleiterfertigung üblichen, alkalischen, H2O2 enthaltenden Reinigungslösungen untersucht. Die Stabilitätsbestimmungen von DEHP, ausgedrückt in Form dessen Halbwertszeit der Zersetzung in den Reinigungslösungen, wurden mit Hilfe der UV/Vis-Spektralphotometrie durchgeführt. Im Zuge der Stabilitätsbestimmungen wurde festgestellt, dass der Komplexbildner DEHP die Reinigungslösungen zwischen drei (im Fall von 1,65/1/5 TPM) bis vier (im Fall von ¼/20 SC1 und von 1,65/1/5 NC) Halbwertszeiten zu stabilisieren vermag. Darüber hinaus wurde festgestellt, dass DEHP die niedrigste Halbwertszeit in Reinigungslösungen mit TMAH als Base und die höchste in solchen mit Cholin besitzt. Die in den ¼/20-SC1-Lösungen, also mit NH3 als Base, nimmt die mittlere Stellung der drei untersuchten Typen von Reinigungslösungen ein. Diese Abstufung der Stabilitäten wird einerseits dem individuellen komplexierenden Potential der einzelnen Basen als auch der von Cholin vermuteten Rolle, in der Reinigungslösung als Opferreduktionsmittel zu wirken und damit die Lebensdauer (Halbwertszeit) des Komplexbildners zu fördern, zugeordnet. Die UV/Vis-Spektralphotometrie hat sich darüber hinaus bei den Untersuchungen als einfach handhabbares und möglicherweise auch leicht automatisierbares Quantifizierungsverfahren zur Implementierung in bereits bestehende Reinigungsanlagen erwiesen.
Folding of RNA molecules into their functional three-dimensional structures is often supported by RNA chaperones, some of which can catalyse the two elementary reactions helix disruption and helix formation. Hfq is one such RNA chaperone, but its strand displacement activity is controversial. Whereas some groups found Hfq to destabilize secondary structures, others did not observe such an activity with their RNA substrates. We studied Hfq’s activities using a set of short RNAs of different thermodynamic stabilities (GC-contents from 4.8% to 61.9%), but constant length. We show that Hfq’s strand displacement as well as its annealing activity are strongly dependent on the substrate’s GC-content. However, this is due to Hfq’s preferred binding of AU-rich sequences and not to the substrate’s thermodynamic stability. Importantly, Hfq catalyses both annealing and strand displacement with comparable rates for different substrates, hinting at RNA strand diffusion and annealing nucleation being rate-limiting for both reactions. Hfq’s strand displacement activity is a result of the thermodynamic destabilization of the RNA through preferred single-strand binding whereas annealing acceleration is independent from Hfq’s thermodynamic influence. Therefore, the two apparently disparate activities annealing acceleration and duplex destabilization are not in energetic conflict with each other.
Transmetallation and oxidative substitution were utilized to prepare examples of group 14, group 6 and group 10 complexes from lithiated or chlorinated 4,4-dimethyl-2-(2-thienyl) oxazoline or its N-alkylated analogs. Two of the product types (2and 5) can be classified as a-thio or remote carbene complexes, depending on the position (3- or 5-) of attachment to the substituted thiophene ring. Spectroscopic measurements as well as crystal and molecular structure determinations clarified the bonding within the new compounds.
The nicotinamide-adenine-dinucleotide (NADH):ubiquinone oxidoreductase (complex I) from the strictly aerobic yeast Y. lipolytica contains at least 26 “accessory” subunits however the significance of most of them remains unknown. The aim of this study was to characterize the role of three accessory subunits of complex I, recently identified: two mitochondrial acyl carrier proteins, ACPM1 and ACPM2 and a sulfurtransferase (st1) subunit. ACPMs are small (approx. 10 kDa) acidic proteins that are homologous to the corresponding central components of prokaryotic fatty acid synthase complexes. Genomic deletions of the two genes ACPM1 and ACPM2 resulted in strains that were not viable or retained only trace amounts of assembled mitochondrial complex I, respectively, as assessed using two-dimensional blue native/sodium dodecyl sulfate polyacrylamide gel electrophoresis (BN/SDS) PAGE. This suggested different functions for the two proteins that despite high similarity could not be complemented by the respective other homolog still expressed in the deletion strains. To test whether complex I was affected by deletion of the ACPM2 gene, its activities in mitochondrial membranes were measured. Consequently, specific inhibitor sensitive dNADH: decylubiquinone (DBQ) oxidoreductase activity was lost completely and a strong decrease in dNADH: hexa-ammine-ruthenium (HAR) oxidoreductase activity was measured. Remarkably, the same phenotypes were observed if just the conserved serine carrying the phosphopantethein moiety was exchanged with alanine. Although this suggested a functional link to the lipid metabolism of mitochondria, using HPLC chromatography no changes in the lipid composition of the organelles were found. Proteomic analysis revealed that both ACPMs were tightly bound to purified mitochondrial complex I. Western blot analysis revealed that the affinity tagged ACPM1 and ACPM2 proteins were exclusively detectable in mitochondrial membranes but not in the mitochondrial matrix as reported for other organisms. Hence it has been concluded that the ACPMs can serve all their possible functions in mitochondrial lipid metabolism and complex I assembly and stabilization as subunits bound to complex I. A protein exhibiting rhodanese (thiosulfate:cyanide sulfurtransferase) activity was found to be associated with homogenous preparation of complex I. From a rhodanese deletion strain, functional complex I that lacked the additional protein but was fully assembled and displayed no functional defects or changes in EPR signature was purified. In contrast to previous suggestions, this indicated that the sulfurtransferase associated with Y. lipolytica complex I is not required for assembly of its iron–sulfur clusters.
