The title compound, C15H14N2O4, has a trans–gauche [O/C/C/C–O/C/C/C] (TG) conformation. The angle between the planes of aromatic rings is 76.4 (3)°. The crystal structure is stabilized by van der Waals interactions and C—H ... O hydrogen bonds. The crystal used was a non-merohedral twin with a fractional contribution of the minor component of 0.443 (5).
The title compound, C14H20O5S·0.5H2O, crystallizes with two organic molecules and a solvent water molecule in the asymmetric unit. In both molecules, the hexapyranosyl rings adopt a slightly distorted chair conformation (5 C 2) with four substituents in equatorial positions and one substituent in an axial position. The main difference between the organic molecules is the dihedral angle between the phenyl ring and the best plane defined by the O—C1—C2—C3 atoms (r.m.s deviations = 0.003 and 0.043 Å) of the hexapyranosyl rings [47.4 (4) and 86.5 (4)°]. In the asymmetric unit, molecules are linked by two strong O—H[cdots, three dots, centered]O hydrogen bonds. In the crystal, the components are linked by a total of 10 distinct O—H[cdots, three dots, centered]O hydrogen bonds, resulting in the formation of a two-dimensional network parallel to the ab plane.
There are two independent molecules in the asymmetric unit of the title compound, C19H24S2. In both molecules, the aliphatic segment of the ligand is in an all-trans conformation: the –S–(CH2)5–S–bridging chain is almost planar (r.m.s. deviation for all non-H atoms = 0.0393 and 0.0796 Å in the two molecules) and maximally extended. Their mean planes form dihedral angles of 4.08 (6)/20.47 (6) and 2.22 (6)/58.19 (6)° with the aromatic rings in the two molecules. The crystal packing is purely governed by weak intermolecular forces.
The title compound, C20H22O4S2, was synthesized by the reaction of 1,4-dibromobutene with methyl thiosalicylate. The aliphatic segment of this ligand is in an all-trans conformation. The bridging chain, –S-(CH2)4-S–, is almost planar (r.m.s. deviation for all non-H atoms: 0.056 Å) and its mean plane forms dihedral angles of 16.60 (7) and 5.80 (2)° with the aromatic rings. In the crystal, the molecules are linked by weak C—H ... O interactions into chains with graph-set notation C(14) along [0 0 1]. The crystal studied was a racemic twin, the ratio of the twin components being 0.27 (9):0.73 (9).
The title compound (also know as azorellanone), C20H32O2, is built up from three fused carbocycles, one five-membered ring and two six-membered rings. The five membered-ring has an envelope conformation, whereas the six-membered rings have a distorted half-chair and a twist–boat conformation. In the crystal, molecules are linked by O—H ... O interactions into zigzag chains with graph-set notation C(8) along [010]. The absolute configuration was assigned on the basis of earlier chemical studies.

The title compound, [systematic name: 5a-acetoxymethyl-3-isopropyl-8-methyl-1,2,3,3a,4,5,5a,6,7,10,10a,10b-dodecahydro-7,10-endo-epidioxycyclohepta[e]indene-3a-carboxylic acid], C22H32O6 (I), is closely related to methyl 5a-acetoxymethyl-3-isopropyl-8-methyl-1,2,3,3a,4,5,5a,6,7,10,10a,10b-dodecahydro-7,10-endo-epidioxycyclohepta[e]indene-3a-carboxylate, (II) [Brito et al., (2008 [triangle]). Acta Cryst. E64, o1209]. There are two molecules in the asymmetric unit, which are linked by two strong intramolecular O—H ... O hydrogen bonds with graph-set motif R 2 2(8). In both (I) and (II), the conformation of the three fused rings are almost identical. The five-membered ring has an envelope conformation, the six-membered ring has a chair conformation and the seven-membered ring has a boat conformation. The most obvious differences between the two compounds is the observed disorder of the acetoxymethyl fragments in both molecules of the asymmetric unit of (I). This disorder is not observed in (II). The crystal structure and the molecular conformation is stabilized by intermolecular C—H ... O hydrogen bonds. The ability to form hydrogen bonds is different in the two compounds. The crystal studied was a non-merohedral twin, the ratio of the twin components being 0.28 (1):0.72 (1)
A novel thiazol‐based ratiometric dye for the detection of local pH values is synthesized, and its properties are characterized by a combination of optical spectroscopy, solid‐state NMR and DNP (dynamic nuclear polarization)‐enhanced solid‐state NMR. This novel dye covers a completely different sensitivity range with its acidic pKa value of 3.5 compared to other established dyes for ratiometric pH detection, such as SNARF. The dye is grafted to the surfaces of mesoporous silica materials, which enables, for the first time, direct in situ measurements of the local pH values in silica mesopores by a simple UV‐vis spectroscopy method. The obtained results, which are in good agreement with previous indirect techniques, indicate a background electrolyte‐dependent pKa shift of at least one pH unit under nanoconfined conditions compared to the pKa of the dye in bulk solution.
The crystal structure of the title salt, [Li(CH3CN)4][B(NCS)4], is composed of discrete cations and anions. Both the Li and B atoms show a tetrahedral coordination by four equal ligands. The acetonitrile and isothiocyanate ligands are linear. The bond angles at the B atom are close to the ideal tetrahedral value [108.92 (18)–109.94 (16)°], but the bond angles at the Li atom show larger deviations [106.15 (17)–113.70 (17)°].
A new polymorph of the title compound, [Pd2(C8H18P)2(C8H19P)2], has been found. It belongs to the triclinic P-1 space group, whereas the known form [Leoni, Sommovigo, Pasquali, Sabatino & Braga (1992 [triangle]), J. Organomet. Chem. 423, 263–270] crystallizes in the monoclinic C2/c space group. The title compound features a dinuclear palladium complex with a planar central Pd2(μ-P)2 core (r.m.s. deviation = 0.003 Å). The Pd—Pd distance of 2.5988 (5) Å is within the range of a PdI—PdI bond. The molecules of both polymorphs are located on a crystallographic centre of inversion. The molecular conformations of the two polymorphs are essentially identical. The crystal packing patterns, on the other hand, are slightly different.
1-(Bromomercurio)ferrocene

The asymmetric unit of the title compound, [Fe(C5H5)(C5H4BrHg)], contains two independent molecules, A and B, in which the Hg-C bond lengths are 2.045 (6) and 2.046 (6) Å, the Hg-Br bond lengths are 2.4511 (9) and 2.4562 (7) Å, and the C-Hg-Br angles are 176.42 (17) and 177.32 (17)°. The two cyclopentadienyl rings of mol-ecule A are eclipsed, while those of mol-ecule B are almost staggered. The HgBr groups are connected by intermolecular Hg⋯Br contacts of 3.3142 (9)-3.4895 (11) Å, forming layers parallel to (001). These layers contain both four-membered (HgBr)2 and eight-membered (HgBr)4 rings. Ferrocene-ferrocene C-H⋯π contacts connect the molecular layers along the c-axis direction.
A mild synthetic method for N-formyl-Met-Leu-Phe-OH (1) is described. After Fmoc solid phase peptide synthesis, on-bead formylation and HPLC purification, more than 30 mg of the fully 13C/15N-labelled tripeptide 1 could be isolated in a typical batch. This peptide can be easily crystallised and is therefore well suited as a standard sample for setting up solid-state NMR experiments.
Ein bekanntes Beispiel für regulatorische RNA-Protein-Wechselwirkung stellt der Komplex der TAR-RNA von HIV-1 und dem viralen Protein Tat dar. Dieser Komplex ist wichtig für die effiziente Transkription des viralen Genoms. Essentiell für die Erkennung der Stem-Loop Struktur ist die Wechselwirkung des Tat-Proteins mit dem aus drei Nukleotiden bestehenden Bulge der TAR-RNA. Das Wissen über die Prinzipien der Erkennung von Tat zu TAR-RNA sollte es möglich machen, spezifische Liganden zu designen, die als antivirale Tat Antagonisten angreifen können. Das Ziel dieser Arbeit war die Synthese von RNA Liganden für die Festphasenpeptidsynthese (FPPS) basierend auf heteroaromatischen Bausteinen. Als Strategie wurde gewählt, die heteroaromatischen Reste über Amid-Bindungen an ein Fmoc geschütztes (2-Aminoethyl)glycin-Rückgrat einzufügen. Die peptidomimetischen Bausteine X konnten in der FPPS zur Synthese von Tripeptiden mit der allgemeinen Struktur Arg-X-Arg und Modifikationen mit Lysin eingesetzt werden. Die Bindungsaffinitäten der Tripeptide und kleinen Moleküle zur TAR-RNA von HIV-1 wurden über einen fluoreszenzbasierten Assay bestimmt. Der Assay verwendet ein doppelt endmarkiertes Tat-Peptid mit Fluorescein und Rhodamin als Farbstoff. Durch Verdrängung des Tat-Peptides durch einen konkurrierenden Liganden kommt es zur Konformationsänderung des Peptides, die zur Löschung der Lichtemmission führt. Dabei zeigen die Tripeptide H2N-(D)Arg-Lactam-(D)Arg-CONH2 (154) und H2N-(D)Arg-Amidin-(D)Arg-CONH2 (158) IC50-Werte von 2-3 mikroM, die auch durch Fluoreszenz Korrelations Spektroskopie (FCS) bestätigt wurden. In massenspektrometrischen Untersuchungen von 158 bzw. Tat-Protein mit TAR-RNA konnten bei beiden Peptiden 1:1 und 1:2-Komplexe beobachtet werden. 158 zeigt antivirale Eigenschaften (IC50 = 10-50 mikroM) in HeLa P4-Zellassays. Durch NMR-Untersuchungen und molekulardynamische Berechnungen war es möglich, eine Konformationsänderung der TAR-RNA durch eine Wechselwirkung mit Guanidinium-Gruppen festzustellen. Ein weiteres Ziel der Arbeit war daher ausgehend von den gewonnenen Daten die Untersuchung des Bindungskonzeptes von Diaminopyrazolen und Indazolen. Diaminopyrazole mit kleinen Resten und Triaminopyrazol übertreffen im protonierten Zustand den dikationischen Liganden Argininamid. In Übereinstimmung mit dem Bindungsmodell verhalten sich protonierte Aminopyrazole wie „Super-Guanidine“ hinsichtlich der TAR-RNA. Das Einfügen des Triaminopyrazols in ein Phenazin-Grundgerüst führt zu Verbindungen, die im protonierten und reduzierten Zustand zusätzliche Wasserstoffbrückenbindungen eingehen können und nicht planar, aber lipophil sind. Der Austausch von Stickstoff zu Sauerstoff im Phenazinring sollte den reduzierten Zustand stabilisieren. Die resultierende Struktur ist jedoch zersetzlich, so dass auf weitere Untersuchungen verzichtet wurde.
7 Zusammenfassung und Ausblick 7.1 Synthese des PNA-Bausteine Primäres Ziel der Diplomarbeit war die Synthese von 6-Aminochinolin-2(1H)-on (3) und dessen Kupplung über die Carbonsäure (4) mit dem PNA-Grundgerüst (6). ... Das Chinolin (3) wurde durch eine dreistufige Synthese aus para-Phenylendiamin (2) erhalten und als Pikrat gefällt. Über eine Ionenaustauschsäule wurde das Hydrochlorid des Chinolins (3b) gewonnen. Durch Charakterisierung des RNA-Liganden mittels FRET konnte ein IC50-Wert des Chinolin Hydrochlorids (3b) von 46 μmol ermittelt werden. Das kupplungsfähige Essigsäurederivat (4) wurde über den tert-Butylesters (87) des Chinolins (3) gewonnen. ... Die literaturbekannte zweistufige Synthese des PNA-Rückgrats (5) geht vom Ethylendiamin ... 7.2 Synthese des PNA-Monomers Das Ziel die Synthese des PNA-Monomers (6), konnte durch Kupplung des Essigsäurederivats (4) und des PNA-Rückgrats (5) mittels DIC/HOBt erreicht werden. ... Das PNA-Monomer sollte in weiterführenden Arbeiten durch kombinatorische Chemie in ein Tripeptid eingebaut werden. Dazu wäre es jedoch notwendig, die tert-Butyl-Schutzgruppe durch Säurezugabe abzuspalten, um die Carbonsäure (88) zu erhalten, die als PNA-Monomer in ein Tripeptid eingebaut werden kann. Es wurde jedoch eine Zersetzung des PNA-Monomers in das Hydrochlorid des Chinolins (3b) und zwei Fragmenten des Fmoc-geschützten Grundgerüsts nachgewiesen. 7.3 Alternative Methoden ... Auf einem anderen Weg konnte das Anhydrid (8) aus Ethylendiamin (9) in vier Syntheseschritten dargestellt werden, das mit dem Chinolin (3) gekuppelt werden konnte. Die resultierende Carbonsäure (10) wurde durch weiterführende Arbeiten in ein Tripeptid eingebaut. Nach der Abspaltung vom Harz und der Entschützung des Tripeptids fand eine intramolekulare Cyclisierung statt, wobei das Chinolin (3) als Abgangsgruppe nachgewiesen wurde. In nachfolgenden Arbeiten sollten die Eigenschaften von Iminochinolin (93) als RNA-Ligand bestimmt werden. Der Syntheseweg sollte über eine dreistufige Synthese vom Chinolin (3) ausgehend zum Iminochinolin (93) führen. Es konnte im Rahmen dieser Arbeit gezeigt werden, dass der erste Schritt, die Darstellung des Chlorochinolins (91), gelingt. 7.4 Ausblick Das erwähnte Iminochinolin (93) sollte in nachfolgenden Arbeiten ebenfalls auf seine Eigenschaft als RNA-Ligand untersucht werden. Die PNA-Monomere ließen sich nur mit geringen Ausbeuten kuppeln. In der Literatur zur Darstellung von PNA-Monomeren werden unterschiedliche Kupplungsreagenzien, wie EDC/HOBt, BOP/HOBt, TOTU oder PyBOP, für die PNA-Grundgerüste angegeben. Um bessere Ausbeuten zu erreichen, ist eine systematische Untersuchung der optimalen Kupplungsbedingungen vorzunehmen. Um die Zersetzung des PNA-Monomers (6) bei der Abspaltung der tert-Butylschutzgruppe zu verhindern, ist nach alternativen PNA-Rückgraden zu suchen. Methyl- oder Ethylester statt tert-Butylester wären durch Verseifung mit Natriumhydroxid in einem Wasser/Dioxan-Gemisch in das jeweilige Carboxylat zu überführen. Aufgrund der niedrigen Ausbeute des Tripeptids (4)-(D)Arg-(D)-Arg-CO-NH2 (96) soll versucht werden die Kupplungsausbeute durch Vermeidung möglicher Nebenreaktionen zu erhöhen. Eine mögliche Nebenreaktion ist die Cyclisierung der zu kuppelnden Carbonsäure (4) mit einer weiteren Carbonsäure (4), die vermieden werden kann durch die Einführung einer Boc-Schutzgruppe am sekundären Amin. Eine Kupplung mit der Boc-geschützten Carbonsäure (97) verspricht bessere Kupplungsausbeuten. ... In vorangegangenen Arbeiten[74][75] konnten das Lactam (98) mittels Heck- oder Suzuki-Reaktion nicht an das Vinylglycinderivat (99) gekuppelt werden. ... Als alternative Kupplungsmethode wird gerade in einer Doktorarbeit die „chiral pool“- Synthese mit Glutaminsäure (101) untersucht, die in die halogenierte Spezies (102) überführt werden soll. Die Kreuzkupplungsreaktion erfolgt durch eine Fe(acac)3 vermittelte Grignard-Reaktion...
Um die Bedeutung bestimmter Neurone oder Klassen von Neuronen innerhalb von Nervensystemen zu untersuchen, sind Methoden, die eine Manipulation der Aktivität der Neurone in vivo erlauben, besonders nützlich. Die bisher zur Verfügung stehenden Methoden haben jedoch Einschränkungen in beispielsweise der Zelltypspezifität, der zeitlichen Präzision, der Reversibilität oder der Anwendbarkeit in frei beweglichen Tieren. Im Rahmen dieser Arbeit wurden optogenetische, d.h. auf der Expression lichtempfindlicher Proteine basierende Methoden entwickelt, um eine präzise Manipulation des Membranpotentials definierter Neurone durch Licht zu ermöglichen. Die Techniken wurden daraufhin zur Untersuchung z.B. der Neurotransmission sowie der Funktion kleiner Netzwerke im Nervensystem des Nematoden Caenorhabditis elegans verwendet. Die zelltypspezifische heterologe Expression des lichtgesteuerten Kationenkanals Channelrhodopsin-2 (ChR2) aus der Grünalge Chlamydomonas reinhardtii ermöglichte es, Muskel- oder Nervenzellen der Nematoden durch blaue Beleuchtung innerhalb weniger Millisekunden zu depolarisieren. Dadurch ließen sich spezifische Verhaltensweisen in frei beweglichen Tieren auslösen. Dieser Ansatz wird in Zukunft die Untersuchung der Bedeutung einzelner Neurone innerhalb ihrer Schaltkreise deutlich erleichtern. So konnte hier gezeigt werden, dass die Photostimulation des propriorezeptiven Neurons DVA eine signifikante Erhöhung der mittleren Körperbiegungswinkel während der sinusförmigen Fortbewegung der Tiere zur Folge hatte. Außerdem wurde versucht, die Anwendbarkeit von ChR2 durch die gezielte Manipulation der subzellulären Lokalisation mittels Fusion mit speziellen Peptiden oder Proteinen zu erhöhen. Des Weiteren wurde eine zur Verwendung von ChR2 analoge Methode zur Hemmung von Neuronen durch Licht entwickelt. Hierfür wurde die lichtgetriebene Cl--Pumpe Halorhodopsin aus Natronomonas pharaonis (NpHR) zelltypspezifisch in C. elegans exprimiert. Durch Photoaktivierung von NpHR mit gelbem Licht war es möglich, Muskelzellen und cholinerge Neurone zu hyperpolarisieren und somit in ihrer Aktivität zu hemmen. Dies führte in frei beweglichen Tieren zu einer augenblicklichen Paralyse verbunden mit einer drastischen Reduktion der Schwimmfrequenz und einer Erhöhung der Körperlänge. Die Aktionsspektren von ChR2 und NpHR sind unterschiedlich genug, um eine unabhängige Photoaktivierung der beiden Proteine mit blauem und gelbem Licht zu ermöglichen. Dadurch konnte die Aktivität von Muskelzellen und cholinergen Neuronen nach Koexpression der beiden Proteine bidirektional kontrolliert werden. Es wurden somit Methoden entwickelt, die in vivo eine zeitlich äußerst präzise Manipulation des Membranpotentials definierter Neurone mit Licht verschiedener Wellenlängen ermöglichen. Die Beobachtung der dadurch induzierten Verhaltensänderungen erlaubt es, zuverlässige Aussagen über die Bedeutung einer Nervenzelle für die Ausprägung eines Verhaltens zu treffen und wird die Erforschung der Nervensysteme von C. elegans und anderen Modellorganismen deutlich vereinfachen. Schließlich wurden in dieser Arbeit optogenetische Methoden zur Untersuchung der synaptischen Übertragung an neuromuskulären Synapsen (neuromuscular junctions, NMJs) von C. elegans entwickelt. Hierfür wurde ChR2 in GABAergen oder cholinergen Neuronen exprimiert, um eine lichtgesteuerte Freisetzung des inhibitorischen Neurotransmitters GABA bzw. des exzitatorischen Neurotransmitters Acetylcholin (ACh) an NMJs zu erreichen. Die Methode wurde OptIoN getauft, ein Akronym für „Optogenetic Investigation of Neurotransmission“, also „optogenetische Untersuchung der Neurotransmission“. Die GABA-Freisetzung hatte ähnlich wie die NpHR-vermittelte Photoinhibition von Muskelzellen eine Reduktion der Schwimmfrequenz und Erhöhung der Körperlänge zur Folge. Die Ausschüttung von ACh verursachte hingegen starke Muskelkontraktionen verbunden mit einer Reduktion der Körperlänge. Die Änderungen der Körperlänge waren bei Mutanten mit verschiedenen Neurotransmissionsdefekten signifikant unterschiedlich im Vergleich zum Wildtyp. Außerdem kam es während längerer Beleuchtungsphasen in Mutanten mit defektem Recycling der synaptischen Vesikel (SV) zu einer verstärkten Abnahme der lichtinduzierten Effekte. OptIoN ermöglicht es dadurch erstmals, die Mechanismen des SV-Recyclings in C. elegans Verhaltensexperimenten zu untersuchen. In elektrophysiologischen Messungen ließen sich durch kurze Lichtpulse wiederholt und mit hoher Frequenz Neurotransmitter-spezifische postsynaptische Ströme evozieren. Diese Ströme waren in Mutanten mit gestörter SVExozytose reduziert und gingen bei wiederholter Stimulation in Mutanten mit defektem SVRecycling schneller zurück. Die Verwendung von OptIoN erleichtert die elektrophysiologische Untersuchung neuronaler Defekte und stellt erstmals eine Möglichkeit dar, Vorgänge der neuronalen Plastizität in dem genetischen Modellsystem C. elegans zu untersuchen. Das Potential von OptIoN zeigte sich unter anderem auch in der Identifizierung eines neuen, über metabotrope GABA-Rezeptoruntereinheiten vermittelten Mechanismus zur heterosynaptischen Hemmung cholinerger Neurone.
The inhalation of particulate matter (PM) in second-hand smoke (SHS) is hazardous to health of smokers and non-smokers. Tobacco strength (amount of tar, nicotine, and carbon monoxide) and different additives might have an effect on the amount of PM. This study aimed to investigate the influence of tobacco strength or additives on PM. Four cigarette types of the brand Marlboro with different strengths and with or without additives were analyzed in comparison to the 3R4F reference cigarette. SHS was generated by an automatic environmental tobacco smoke emitter (AETSE) in an enclosed space with a volume of 2.88 m³. PM concentrations (PM10, PM2.5, PM1) were measured with a laser aerosol spectrometer followed by statistical analysis. The two strongest Marlboro brands (Red and Red without additives) showed the highest PM concentrations of all tested cigarettes. The measured mean concentrations Cmean of PM10 increased up to 1458 µg/m³ for the Marlboro Red without additives (PM2.5: 1452 µg/m³, PM1: 1263 µg/m³). The similarly strong Marlboro Red showed very similar PM values. The second strongest type Marlboro Gold showed 36% (PM10, PM2.5) and 32% (PM1) lower values, respectively. The “lightest” type Marlboro Silver Blue showed 54% (PM10, PM2.5) or 50% (PM1) lower PM values. The results indicate that the lower the tar, nicotine, and carbon monoxide amounts, as well as the longer the cigarette filter, the lower are the PM levels. An influence of additives could not be determined.
Children are commonly exposed to second-hand smoke (SHS) in the domestic environment or inside vehicles of smokers. Unfortunately, prenatal tobacco smoke (PTS) exposure is still common, too. SHS is hazardous to the health of smokers and non-smokers, but especially to that of children. SHS and PTS increase the risk for children to develop cancers and can trigger or worsen asthma and allergies, modulate the immune status, and is harmful to lung, heart and blood vessels. Smoking during pregnancy can cause pregnancy complications and poor birth outcomes as well as changes in the development of the foetus. Lately, some of the molecular and genetic mechanisms that cause adverse health effects in children have been identified. In this review, some of the current insights are discussed. In this regard, it has been found in children that SHS and PTS exposure is associated with changes in levels of enzymes, hormones, and expression of genes, micro RNAs, and proteins. PTS and SHS exposure are major elicitors of mechanisms of oxidative stress. Genetic predisposition can compound the health effects of PTS and SHS exposure. Epigenetic effects might influence in utero gene expression and disease susceptibility. Hence, the limitation of domestic and public exposure to SHS as well as PTS exposure has to be in the focus of policymakers and the public in order to save the health of children at an early age. Global substantial smoke-free policies, health communication campaigns, and behavioural interventions are useful and should be mandatory.
Objective: Inhaled particulate matter (PM) in secondhand smoke (SHS) is deleterious for smokers and non-smokers. Different additives in cigarettes might effect the amount of PM. This study aimed to assess the influence of additives on the PM emissions from different cigarette types in SHS.
Design: An experimental study of PM measuring in SHS of cigarettes without exposition of any person.
Method: The concentrations of PM (PM10, PM2.5 and PM1) in SHS of four different types of cigarettes of the brand Lucky Strike, two types with additives (Original Red, Original Blue) and two types without additives (Straight Red, Straight Blue), in comparison to the reference cigarette 3R4F were analysed. An automatic environmental tobacco smoke emitter generated SHS in an enclosed space with a volume of 2.88 m3. PM was measured with a laser aerosol spectrometer (Grimm model 1.109). Afterwards, the measuring values of the four Lucky Strike brands and the reference cigarette were statistically evaluated and visualised.
Results: Lucky Strike Straight Blue, a cigarette type without additives and lower tar amount, showed 10% to 25% lower PM mean values compared with the other tested Lucky Strike products, but 21% (PM1) respectively 27% (PM2.5,PM10) higher mean values than the reference cigarette. The PM mean of all measured smoke-free baseline values (clean air) was 1.6 µg/m³. It increased up to about 1800 µg/m³ for the reference cigarette and up to about 3070 µg/m³ for the Lucky Strike Original Blue.
Conclusions: The findings of this study show the massive increase of PM amount by smoking cigarettes in enclosed spaces and suggest that additives in tobacco products increase the PM amount in SHS. For validation, further comparative studies are necessary focusing on the comparison of the PM concentration of cigarettes with and without additives.
Implications: Due to the exposure to SHS, 890 000 people die each year worldwide. PM in SHS endangers the health of both non-smokers and smokers. This study considers the effect of additives like aromatics and humectant agents in cigarettes on PM in SHS. Do additives in tobacco products increase the amount of PM?
The influence of temperatur and pressure on the fluorescence quantum yield of N-methylacridone (9,10-dihydro-9-oxo-10-methyl-acridine) in toluene in the range of 283-313 K and 1 bar to 2.5 kbar, respectively, has been investigated. Treatment of the data in terms of the Eyring transition-state theory leads to a consistent interpretation of the observed effect. The unusually large increase of the quantum yield with increasing pressure is attributed to a positive volume of activation, ⊿V≠, for the thermally activated S1-T2 intersystem crossing which is known to be the only deactivation process (of the Si-state) competing with fluorescence. Comparison of the values for ⊿H≠, the activation enthalpy of this process, determined at various pressures, indicates a decrease in ⊿H≠ at elevated pressures. Since ⊿H≠ can be associated with the S1-T2 energy gap involved in intersystem crossing, this result further confirms the conclusion that the change in Franck-Condon factors alone cannot account for the decrease in the intersystem crossing rate with increasing pressure.
The immune system makes use of major histocompatibility complex class I (MHC I) molecules to present peptides to other immune cells, which can evoke an immune response. Within this process of antigen presentation, the MHC I peptide loading complex, consisting of a transporter associated with antigen processing TAP, MHC I, and chaperones, is key to the initiation of immune response by shuttling peptides from the cytosol into the ER lumen. However, it is still enigmatic how the flux of antigens is precisely coordinated in time and space, limiting our understanding of antigen presentation pathways. Here, we report on the development of a synthetic viral TAP inhibitor that can be cleaved by light. This photo-conditional inhibitor shows temporal blockade of TAP-mediated antigen translocation, which is unleashed upon illumination. The recovery of TAP activity was monitored at single-cell resolution both in human immune cell lines and primary cells. The development of a photo-conditional TAP inhibitor thus expands the repertoire of chemical intervention tools for immunological processes.
A great challenge in life sciences remains the site-specific modification of proteins with minimal perturbation for in vitro as well as in vivo studies. Therefore, different chemoselective reactions and semi-synthetic techniques such as native chemical ligation or intein-mediated protein splicing have been established. They enable a site-specific incorporation of chemical reporters into proteins, such as organic fluorophores or unnatural amino acids. In this PhD Thesis, protein trans-splicing was guided by minimal high-affinity interaction pairs to trace proteins in mammalian cells. In addition, the temporal modulation of cellular processes by photo-cleavable viral immune evasins was achieved.
Protein trans-splicing mediated by split inteins is a powerful technique for site-specific and 'traceless' protein modifications. Despite recent developments there is still an urgent need for ultra-small high-affinity intein tags for in vitro and in vivo approaches. So far, only a very few in-cell applications of protein trans-splicing are reported, all limited to C-terminal protein modifications. Here, a strategy for covalent N-terminal intein-mediated protein labeling at sub-nanomolar probe concentrations was developed. Combined with the minimalistic Ni-trisNTA/His-tag interaction pair, the affinity between the intein fragments was increased 50-fold (KD ~ 10 nM). Site-specific and efficient 'traceless' protein modification by high-affinity trans-splicing is demonstrated at nanomolar concentrations in mammalian cells.
High background originating from non-reacted, 'always-on' fluorescent probes still is a crucial issue in life sciences. Covalent labeling approaches with simultaneous activation of fluorescence are advantageous to increase sensitivity and to reduce background signal. Therefore, high-affinity protein trans-splicing was combined with fluorophore/quencher pairs for online detection of covalent N-terminal protein labeling in cellular environments. Substantial fluorescence enhancement at nanomolar probe concentrations was achieved. This ultra-small fluorogenic high-affinity split intein system is an unprecedented example for real-time monitoring of the trans-splicing reaction in cell-like environments as well as for protein labeling with fluorogenic probes at nanomolar concentrations.
To extend the field of chemical immunology and to address spatiotemporal aspects in adaptive immune response, new tools to control antigen processing are required. Therefore, synthetic photo-conditional viral immune evasins were designed to modulate antigen processing on demand. By using light, the time and dose controlled antigen translocation by the transporter associated with antigen processing (TAP) was triggered with response in the second regime. Peptide delivery and loading by the peptide-loading complex (PLC) was rendered inactive, whereas blocking was abolished in a light-controlled fashion to inactivate the synthetic viral immune evasin ICP47 along with simultaneous activation of the antigen presentation pathway. Lightresponsive peptide translocation by the TAP complex was assayed in vitro by utilizing microsomes isolated from professional antigen presenting B-cell lymphomas (Raji). To extend these studies, suppression and photo-controlled rescue of antigen presentation was examined at single-cell resolution in human primary immune cells.
Native chemical ligation interconnects peptide chemistry with recombinantly expressed proteins. This technique was applied to generate the semi-synthetic full-length ICP47. Although this approach was realized, the low product yield was not sufficient for further functional studies. Therefore, full-length ICP47 was consecutively generated by utilizing a full synthetic four-fragment ligation approach. However, this synthetic viral immune evasin was not able to block peptide translocation in a robust way.
Ziel dieser Arbeit war die Bestimmung und Verfeinerung der Lösungsstruktur der rekombinanten pI=5,1 Isoform des H-FABPs aus Rinderherz. Dabei wurde von Proben mit einem Fettsäuregemisch ausgegangen und somit die HOLO–Form des Proteins untersucht. Der Einsatz von mehrdimensionaler NMR–Spektroskopie zur Bestimmung von Abstandsrandbedingungen, stereospezi…schen Zuordnungen und Diederwinkeln aus 3J–Kopplungskonstanten ermöglichte die Verwendung von 1877 Abstandsrandbedingungen, 52 stereospezifischen Zuordnungen und 81 Diederwinkelbeschränkungen. Die Struktur basiert im Schnitt auf 15 geometrischen Beschränkungen je Aminosäurerest. Mit einem mittleren globalen RMSD–Wert der Rückgratatome von 1,07+-0,15Å und den relativ niedrigen berechneten Energiewerten sind die Ansprüche an eine hochaufgelöste Struktur erfüllt. Vergleiche mehrerer struktureller Kriterienmit 160 Proteinkristallstrukturen haben ergeben, daß die hier bestimmte Lösungsstruktur in ihrer Güte einer Kristallstruktur mit einer Auflösung von etwa 2,4 Å entspricht. Die Molekulardynamiksimulation der energetisch niedrigsten Lösungsstruktur in einer Wassersimulation führte anfangs zu einer raschen Konformationsänderung der Struktur. Diese war gekennzeichnet durch eine Vergrößerung des als Eingangsportal für den Fettsäureliganden postulierten Bereiches. Zugleich wurden zwei weitere kleine Öffnungen erkennbar, welche als Austrittsöffnungen für Wassermoleküle dienen könnten.
In dieser Arbeit wurde eine Geometrieuntersuchung am FABP-Molekülmodell durchgeführt. Um 3J-Kopplungsinformation für die Diederwinkelanalyse zu bestimmen, wurden J-modulierte [15N,1H]-COSY-Experimente durchgeführt. Mit Hilfe numerischer Anpassungsroutinen wurden die Signalintensitäten quantitativ ausgewertet, um sehr genaue 3JHN,H-alpha-Kopplungskonstanten zu erhalten. Diese Kopplungskonstanten wurden in Kraftfeldrechnungen als experimentelle Randbedingungen für die Phi-Diederwinkel des Proteinrückgrates berücksichtigt. Dadurch ist eine Aussage über die Winkelverteilung und über die zeitlich gemittelten Kopplungseffekte im Proteinmodell möglich. Die aus der Molekulardynamiksimulation bestimmten 3JHN,H-alpha-Kopplungskonstanten wurden mit den experimentellen verglichen. Die Analyse ergab, daß die den beta-Faltblättern zugewandten Ränder der alpha-Helixbereiche sowie zwei der zehn beta-Faltblattbereiche sehr flexible Teilabschnitte aufweisen. Diese Arbeit konnte somit die Überlegungen stützen, die vor allem den flexiblen Teilabschnitt zwischen der alpha-II-Helix und dem beta-B-Faltblattbereich für die Funktion des Proteins verantwortlich machen.
Small interfering RNAs (siRNAs) are now established as the preferred tool to inhibit gene function in mammalian cells yet trigger unintended gene silencing due to their inherent miRNA-like behavior. Such off-target effects are primarily mediated by the sequence-specific interaction between the siRNA seed regions (position 2–8 of either siRNA strand counting from the 5'-end) and complementary sequences in the 3'UTR of (off-) targets. It was previously shown that chemical modification of siRNAs can reduce off-targeting but only very few modifications have been tested leaving more to be identified. Here we developed a luciferase reporter-based assay suitable to monitor siRNA off-targeting in a high throughput manner using stable cell lines. We investigated the impact of chemically modifying single nucleotide positions within the siRNA seed on siRNA function and off-targeting using 10 different types of chemical modifications, three different target sequences and three siRNA concentrations. We found several differently modified siRNAs to exercise reduced off-targeting yet incorporation of the strongly destabilizing unlocked nucleic acid (UNA) modification into position 7 of the siRNA most potently reduced off-targeting for all tested sequences. Notably, such position-specific destabilization of siRNA–target interactions did not significantly reduce siRNA potency and is therefore well suited for future siRNA designs especially for applications in vivo where siRNA concentrations, expectedly, will be low.
The use of chemically synthesized short interfering RNAs (siRNAs) is currently the method of choice to manipulate gene expression in mammalian cell culture, yet improvements of siRNA design is expectably required for successful application in vivo. Several studies have aimed at improving siRNA performance through the introduction of chemical modifications but a direct comparison of these results is difficult. We have directly compared the effect of 21 types of chemical modifications on siRNA activity and toxicity in a total of 2160 siRNA duplexes. We demonstrate that siRNA activity is primarily enhanced by favouring the incorporation of the intended antisense strand during RNA-induced silencing complex (RISC) loading by modulation of siRNA thermodynamic asymmetry and engineering of siRNA 3-overhangs. Collectively, our results provide unique insights into the tolerance for chemical modifications and provide a simple guide to successful chemical modification of siRNAs with improved activity, stability and low toxicity.
Starkes Übergewicht und eine damit einhergehende Hypertrophie von Geweben aber auch des Herz-Kreislauf-Systems führen zu einer Reihe von Folgeerkrankungen wie z. B. Diabetes mellitus Typ 2 oder auch Arteriosklerose. Während im Fettgewebe freie Fettsäuren, die von Makrophagen aufgenommen werden, eine entscheidende Rolle spielen, scheint in der Pathogenese von Arteriosklerose die Aufnahme von Fettsäuren aus Lipoproteinpartikeln durch Makrophagen von großer Wichtigkeit zu sein. Ein weiterer Faktor, der durch freie Fettsäuren ausgelöst wird ist ER-Stress. Makrophagen, die zu Triglycerid (TG) reichen Schaumzellen geworden sind, akkumulieren in arteriosklerotischen Läsionen. Der Lipidmetabolismus von Makrophagen wird transkriptionell u.a. durch den Transkriptionsfaktor PPARγ (Peroxisomproliferator aktivierter Rezeptor γ) reguliert. Sein Zielgen FABP4 (Fettsäuren bindendes Protein 4) beschleunigt die Entwicklung von Arteriosklerose in Mausmodellen. Da die Expression von PPARγ und FABP4 in IL 4- (Interleukin-4) polarisierten Makrophagen induziert wird, sollte die Rolle von FABP4 in humanen, mit IL 4 polarisierten Makrophagen untersucht werden. Hierfür wurden primäre humane Monozyten in Anwesenheit von LPS/IFNγ (Lipopolysaccharid/Interferon γ) bzw. IL 4 zu Makrophagen differenziert. Es zeigte sich, dass in LPS/IFNγ stimulierten Makrophagen PPARγ und dessen Zielgene nicht exprimiert wurden. Dagegen waren sie bei unstimulierten Makrophagen bei IL 4 stimulierten Makrophagen deutlich erhöht. Dies spiegelte sich auch in einer erhöhten Aufnahme von Triglyceriden aus VLDL-Partikeln (Lipoproteinpartikel sehr niedriger Dichte) wider. IL 4 induzierte also einen Fettsäuren akkumulierenden Phänotyp. Durch einen PPAR-Luciferase-Reporter-Test wurde untersucht, ob FABP4 für die Aktivierung von PPARγ nötig war. Dies konnte bestätigt werden, da PPARγ durch seinen Liganden Linolsäure nur in Anwesenheit von FABP4 aktiviert werden konnte. Diese Aktivierung konnte zusätzlich durch den FABP4-Inhibitor HTS01037 verhindert werden. Nun sollte der Einfluss von FABP4 auf die PPARγ-abhängige Genexpression untersucht werden. Hierfür wurde FABP4 während der Differenzierung mit den beiden Inhibitoren HTS01037 oder BMS309403 in IL 4 stimulierten Makrophagen inhibiert. Durch die Inhibition von FABP4 sank die Expression von FABP4 und LPL (Lipoproteinlipase), während die von PPARγ unverändert blieb. Die LPL spielt eine entscheidende Rolle in der Aufnahme von Lipiden aus VLDL-Partikeln und trägt somit zur TG-reichen Schaumzellbildung bei. Die verminderte Expression von LPL spiegelte sich in einer verminderten Lipidaufnahme aus VLDL-Partikeln wider. Gleichzeitig wurde durch die FABP4-Inhibition die Entzündungsantwort der Makrophagen auf VLDL-Partikel abgeschwächt. IL 4 induziert also LPL, indem es PPARγ aktiviert. FABP4 unterstützt hierbei die Aktivierung von PPARγ. Durch die Inhibition kann die LPL-Expression vermindert werden, was die TG-reiche Schaumzellbildung und die Entzündungsreaktion in einem VLDL-reichen Umfeld vermindert und eine neue Therapiemöglichkeit von Arteriosklerose eröffnet. Im Fettgewebe kommt bei starkem Übergewicht, bedingt durch die erhöhte Konzentration an freien Fettsäuren und Hypoxie, zu einer leichten Entzündungsreaktion. Diese Entzündungsreaktion wurde durch eine Stimulation mit Palmitat unter Hypoxie (1 % O2) nachgebildet. Überstände von Makrophagen nach dieser Stimulation (MCM) wurden auf primäre humane Adipozyten übertragen. Diese Überstände konnten zwar keine Insulinresistenz in Adipozyten auslösen, induzierten jedoch eine Entzündungsreaktion. Diese zeigte sich in einer erhöhten Expression der proentzündlichen Zytokine CCL2 (CC-Chemokin-Ligand-2) und IL 6. Gleichzeitig wurde die Expression des antientzündlichen Zytokins Adiponectin vermindert. Der Transfer von MCM ist also ein Modell für die Entstehung der Insulinresistenz in einem frühen Stadium. Beim Versuch, die entzündungsfördernde Fähigkeit des MCMs zu verhindern, wurde AMPK mit verschiedenen Aktivatoren stimuliert. Es zeigte sich, dass der AMPK-Aktivator AICAR (5-Aminoimidazol-4-carboxamidribonukleotid) die Entzündungsantwort und den ER-Stress von mit Hypoxie und Palmitat stimulierten Makrophagen deutlich reduzierte. Der starke Effekt auf den ER-Stress konnte auch mit anderen ER-Stress-Auslösern wie Thapsigargin oder Tunicamycin nachvollzogen werden. Da AICAR ein AMPK-Aktivator ist, wurden typische Effekte der AMPK-Aktvierung wie reduzierte Proteinexpression, verstärkte Sirtuin-1-Aktivierung und Steigerung der Fettsäurenoxidation mittels Inhibitoren verhindert. Dies hatte keinen Einfluss auf die Wirkung von AICAR. Ebenso wurde untersucht, ob AICAR in die Zelle aufgenommen werden musste und ob es zu seiner phosphorylierten Form ZMP umgewandelt werden musste. Durch den Inhibitor ABT 702 kann die Adenosinkinase inhibiert werden, welche die Phosphorylierung katalysiert. Es zeigte sich, dass die Phosphorylierung von AICAR zu ZMP nicht erforderlich war, damit AICAR die ER-Stress-Antwort hemmen konnte. AICAR und nicht ZMP wirkte gegen den ER-Stress. Da durch das fehlende ZMP die AMPK nicht aktiviert wurde, war das ein weiteres Zeichen, dass AICAR AMPK-unabhängig wirkte. Dies konnte durch einen AMPK-Knockdown bestätigt werden. Durch einen Knockdown verschiedener Adenosintransporter konnte gezeigt werden, dass SLC28A3 (Soluttransporterfamlie 28 Typ A3) verantwortlich für die Aufnahme von AICAR in primäre humane Makrophagen war. Es konnte demnach gezeigt werden, dass AICAR den ER-Stress in primären humanen Makrophagen in einem von AMPK unabhängigen Mechanismus vermindert. Dafür wird es mittels SLC28A3 in die Zelle aufgenommen und wirkt als AICAR und nicht als ZMP. Diese Erkenntnisse stellen eine interessante, neue therapeutische Möglichkeit im Feld von Arteriosklerose und Diabetes dar.
Im Rahmen dieser Doktorarbeit wurden modifizierte Nukleoside synthetisiert, um ihren Einfluss auf die Stabilität von RNA-Duplexen zu untersuchen. Bei den fluorierten Benzimidazol-Nukleosidanaloga handelt es sich um universelle Basen, die bei der Basenpaarung nicht zwischen den vier natürlichen Nukleosiden unterscheiden können. Die dabei auftretende Destabilisierung der RNA-Duplexe sollte durch die Änderung physikalisch-chemischer Eigenschaften vermindert werden. Durch die Synthese der fluorierten Indol-Nukleosidanaloga mit denselben Fluoratompositionen sollte nachgewiesen werden, welche Rolle ein ausfallendes Stickstoffatom im Fünfring-System spielt. Weitere Untersuchungen wurden so entwickelt, dass die zwei spC-F in 4,6DFBI wie auch in 4,6DFI mit Stickstoffatomen getauscht wurden. So wurde noch eine neue Serie Nukleosidanaloga synthetisiert (Abbildung 9.2). Schließlich wurde noch 1-Desoxy-D-ribofuranose AS als absischer Baustein synthetisiert. Die Synthese der Indol- und 9-Deazapurin-Nukleosidanaloga wurde über eine Glycsilierungsreaktion mit geeignet geschützter Deoxyribose durchgeführt. Dies wurde über vier Stufen, ohne Aufreinigung, aus Deoxyribose synthetisiert. Die entsprechenden Deoxy-nukleoside wurden danach in fünf Schritten zu Ribo-nukleosiden transformiert. Nach der Entschützung von Toluoyl-Gruppen wurden die 5´- und 3´-OH Gruppen sukzessiv geschützt. Nach simultaner 5´-OH Entschützung und 3´-OMs Eliminierung, wurden die gewünschten Ribonukleoside durch katalytische Dihydroxilierung erhalten. Die Darstellung der Verbindung 7NP erfolgte über die Silyl-Hilbert-Johnson-Reaktion. Der abasische Baustein AS wurde ausgehend von 2,3,5-Tri-O-benzyl-ribofuranose durch Dehydroxylierung und anschließende Entschützung erreicht. Von allen Nukleosiden gelang es Kristalle aus Wasser oder Methanol zu erhalten und röntgenkristallographisch zu untersuchen. Die Kristallpackungen zeigten eine sehr interessante Anordnung der Moleküle. Alle Fluorindol-Nukleoside mit Ausnahme von 7-N-Purin-Nukleosid 7NP zeigten nicht die für aromatische Systeme normale Fischgräten-Struktur, sondern eine Anordnung, in der die Moleküle gegenüberliegen. Die Kristallpackung besteht abwechselnd aus hydrophilen und lipophilen Schichten. Die hydrophilen Schichten bestehen aus den Zuckeruntereinheiten und die lipophilen aus den Fluoraromaten. Die Zucker sind durch Wasserstoffbrücken miteinander verbunden. Für die Orientierung der Moleküle zueinander sind aber die Fluoratome verantwortlich. In der Kristallpackung von 7-Fluorindol-Nukleosid 7FI kann ein Fluor-Wasserstoff-Abstand von nur 230 pm detektiert werden. Dies ist deutlich kürzer als die Summe der van-der-Waals Radien von Fluor und Wasserstoff von 2,55 Å. Der Abstand wird zwischen dem Fluor des einen Nukleosids und einem Wasserstoff eines gegenüberliegenden Nukleosids gemessen. Der Abstand von 2,30 Å ist einer der kürzesten jemals in Kristallen gemessenen F-H Abstände des Typs Csp²-F...H-Csp². Bedingt durch diesen kurzen Abstand kann von einer F...H Wasserstoffbrücke gesprochen werden. Auf der anderen Seite in der Kristallstruktur von 4-Fluorindol-Nukleosid 4FI konnte ein F-H Abstand von 2,69 Å nachgewiesen werden, welcher deutlich länger als die Summe der van-der-Waals Radien von Fluor und Wasserstoff ist. Die Nukleoside wurden auf ihre Lipophilie hin untersucht. Zu diesem Zweck wurden Octanol-Wasser Verteilungskoeffizienten der Nukleoside gemessen. Die fluorierten Nukleoside zeigten im Gegensatz zu den nichtfluorierten Nukleosiden eine deutlich größere Lipophilie. Nach Umsetzung der Nukleoside zu den Phosphoramiditen konnten diese kupplungsfähigen Monomere in den RNA-Festphasensynthesen eingesetzt und in RNA 12mere eingebaut werden. Um den Einfluss der aromatischen Fluorosubstitutionen auf die thermodynamische Stabilität von RNA-Duplexen zu untersuchen, wurden UV/VIS- und CD- spektroskopische Messungen an monomodifizierten RNA 12meren durchgeführt. Aus den erhaltenen Schmelzkurven wurden die Schmelzpunkte bestimmt (Abbildung 9.3) und die thermodynamischen Daten ausgerechnet. Die Anwendung hydrophober, Fluorsubstituierter Nukleobasen führte im Fall der fluorierten Indol-Nukleoside zu Destabilisierung im Vergleich mit natürlichen Basenpaaren. Aus den folgenden Resultaten lässt sich zusammenfassen: 1. Position der Fluoratom in fluorierten Indole spielt eine wesentliche Rolle für die Stabilität des RNA-Duplex 2. 6FI bildet die stabilste Basenpaaren mit natürlichen Basen. 3. Basenpaarung von 4FI trägt eine deutlich höhere Destabilisierung. Für diese Modifikation wurden auch die längsten Abstandwerte zwischen C-F…H in der Kristallpackung gemessen. (Die Vermutung liegt nahe, dass diese Base sich außerhalb des Duplex befindet). 4. Alle Fluorindol-Basenanaloga zeigen die Tendenz zur Paarung mit Adenosin. 5. Bei 4,6DFI handelt sich um universelle Base. Um noch weniger destabilisierende universelle Basen zu finden, wurde das Forschungsfeld mit Methoden aus dem Bereich der strukturellen Bioinformatik, Molekül-dynamiksimulationen und freie Energie-Rechnungen ausgeweitet. Resultierende Simulationen führten zu zwei neuen Basen: 7NP als Analogon zu 4,6DFBI und 9DP als Analogon zu 4,6DFI (siehe Kapitel 8). Theoretische Rechnungen ließen sich bestätigen durch experimentelle Ergebnisse Die so entstandene Serie von Purin-Basenanaloga hat uns gezeigt, dass der Austausch von Fluoratomen durch Stickstoffatome stabilisierende Effekte bringt. Die chemischen Änderungen beeinflussen die physikalischen Eigenschaften, welche dadurch Stabilisierung oder Destabilisierung des RNA-Duuplex dirigieren. In Abbildung 9.5 befinden sich ausgerechnete Dipolmomente. Somit können wir für diese Serie folgendes resümieren: * 4,6FI als universelles Base Analogon zu 4,6DFBI zeigt geringere destabilisierende Effekte auf den 12mer RNA-Duplex. * Umtausch von Fluoratomen in den beiden Basen (4,6DFI und 4,6DFBI) resultiert in deutlich besserer Basenpaarung. * Auserrechnete thermodynamische Parametern (von gemessenen Tm-Werten) wurde ersichtlicht, dass höhere Tm-Werte durch geringere Destabilisierung aus Solvatation resultieren, nicht aus erhöhten Stacking Effekten des RNA-Duplex.
Orientation-selective DEER (Double Electron-Electron Resonance) measurements were conducted on a series of rigid and flexible molecules containing Cu(II) ions. A system with two rigidly held Cu(II) ions was afforded by the protein homo-dimer of copper amine oxidase from Arthrobacter globiformis. This system provided experimental DEER data between two Cu(II) ions with a well-defined distance and relative orientation to assess the accuracy of the methodology. Evaluation of orientation-selective DEER (os DEER) on systems with limited flexibility was probed using a series of porphyrin-based Cu(II)–nitroxide and Cu(II)–Cu(II) model systems of well-defined lengths synthesized for this project. Density functional theory was employed to generate molecular models of the conformers for each porphyrin-based Cu(II) dimer studied. Excellent agreement was found between DEER traces simulated using these computed conformers and the experimental data. The performance of different parameterised structural models in simulating the experimental DEER data was also investigated. The results of this analysis demonstrate the degree to which the DEER data define the relative orientation of the two Cu(II) ions and highlight the need to choose a parameterised model that captures the essential features of the flexibility (rotational freedom) of the system being studied.
The core of photosystem I (PS1) is composed of the two related integral membrane polypeptides, PsaA and PsaB, which bind two symmetrical branches of cofactors, each consisting of two chlorophylls and a phylloquinone, that potentially link the primary electron donor and the tertiary acceptor. In an effort to identify amino acid residues near the phylloquinone binding sites, all tryptophans and histidines that are conserved between PsaA and PsaB in the region of the 10th and 11th transmembrane alpha-helices were mutated in Chlamydomonas reinhardtii. The mutant PS1 reaction centers appear to assemble normally and possess photochemical activity. An electron paramagnetic resonance (EPR) signal attributed to the phylloquinone anion radical (A(1)(-)) can be observed either transiently or after illumination of reaction centers with pre-reduced iron-sulfur clusters. Mutation of PsaA-Trp(693) to Phe resulted in an inability to photo-accumulate A(1)(-), whereas mutation of the analogous tryptophan in PsaB (PsaB-Trp(673)) did not produce this effect. The PsaA-W693F mutation also produced spectral changes in the time-resolved EPR spectrum of the P(700)(+) A(1)(-) radical pair, whereas the analogous mutation in PsaB had no observable effect. These observations indicate that the A(1)(-) phylloquinone radical observed by EPR occupies the phylloquinone-binding site containing PsaA-Trp(693). However, mutation of either tryptophan accelerated charge recombination from the terminal Fe-S clusters.
PPARs gehören wie die Steroidhormon-Rezeptoren (z. B.Glucocorticoid-, Estrogen- oder Testosteron-Rezeptoren) zur Superfamilie der nukleären Rezeptoren. Es existieren drei PPAR-Subtypen, die als PPARalpha, PPARbeta/delta und PPARgamma bezeichnet werden und von drei verschiedenen Genen kodiert werden. Fibrate sind PPARalpha-Agonisten, welche die Plasma-Triglyceridspiegel reduzieren und gleichzeitig eine moderate Steigerung des HDL-Cholesterols bewirken. Thiazolidindione (Glitazone) sind PPARgamma-Agonisten, die bei Typ-2-Diabetes-mellitus indiziert sind und als Insulinsensitizer wirken. Duale PPARalpha/gamma-Agonisten stellen eine neue Klasse von Arzneistoffen dar, die zukünftig zur Behandlung von Typ-2-Diabetikern mit gestörtem Lipidprofil eingesetzt werden könnten. Im Rahmen der vorliegenden Dissertation wurde eine Leitstrukturoptimierung des selektiven PPARalpha-Agonisten Pirinixinsäure (WY 14643) durchgeführt. Die pharmakologische In-vitro-Charakterisierung der Substanzen erfolgte mit Hilfe subtypspezifischer Reportergen-Assays. Zusätzlich wurde gezeigt130, dass Chinolinderivate der Pirinixinsäure 5-LOX-inhibierende Eigenschaften in polymorphnukleären Leukozyten zeigen. Zunächst wurde eine geeignete Synthesestrategie zur Darstellung von Pirinixinsäurederivaten etabliert, was zur Charakterisierung und Identifizierung einer Serie von potenten dualen PPARalpha/gamma-Agonisten und 5-LOX-Inhibitoren führte. Die Synthesestrategie zur Darstellung von sowohl im Arylamino-Bereich (W) als auch in alpha- Position (R) modifizierten Derivaten der Pirinixinsäure bestand in einer vierstufigen Reaktionsfolge (I–IV; siehe Abb.). ... Die PPAR-Modulatoren (Carbonsäuren) 4, 8, 14, 32–38, 41 und 42 wurden in-vitropharmakologisch unter Anwendung subtypselektiver Reportergen-Assays (hPPARalpha, beta, gamma) charakterisiert. Die 5-LOX-Modulatoren (Carbonsäureester) 3, 7, 8, 13, 14, 24–29, 31–42 und 50 wurden in-vitro-pharmakologisch unter Anwendung des 5-LOX-Standardassays in intakten PMNL (polymorphnukleäre neutrophile Leukozyten) evaluiert. Die vorliegende Untersuchung deckte auf, dass der Ersatz des 2,3-Dimethylanilin-Strukturelements der Pirinixinsäure durch 6-Aminochinolin zu einem Gesamtverlust des PPARalpha/gamma-Agonismus führt. Durch Strukturmodifikationen im Arylamino-Bereich der Leitstruktur WY 14643 mit für 5-LOX-Non-Redoxinhibitoren typischen Pharmakophorresten, Tetrahydropyran-4-yl (Substanzen 13–14) und Methoxychinolin-2-yl (Substanz 43), konnte eine signifikante Steigerung des 5-LOX-inhibitorischen Potenzials erzielt werden [IC50 7 mikro M (13) bzw. 4 mikro M (43)]. Die alpha-alkylsubstituierten Carbonsäurederivate 33–38 zeigten sowohl am hPPARalpha als auch am hPPARgamma eine mit der aliphatischen Kettenlänge steigende Potenz. Am hPPARalpha erwiesen sich das alpha-Hexyl-Derivat 35 und das alpha-Butyl-Derivat 34 mit EC50-Werten von 1,9 mikro M (35) bzw. 11,5 mikro M (34) entsprechend einer Steigerung der Aktivität gegenüber WY 14643 um den Faktor 20 (35) bzw. den Faktor 4 (34) als besonders potent. Im Falle des hPPARgamma zeigten ebenso das alpha-butylsubstituierte Derivat 34 und der alpha-hexylsubstituierte Ligand 35 die höchste Aktivität mit EC50-Werten von 8,7 mikro M (34) bzw. 5,8 mikro M (35) und einer Steigerung der Aktivität gegenüber WY 14643 von Faktor 6 (35) bzw. Faktor 9 (34). Die Einführung von alpha-Alkylsubstituenten in das Grundgerüst der Pirinixinsäure führt möglicherweise zum Besetzen der linken proximalen Bindungstasche und somit zu einer im Vergleich zur Leitstruktur WY14643 stärkeren Bindung in die Ligandenbindungstasche des Rezeptors. Die Besetzung dieser proximalen Bindungstasche entscheidet jedoch nicht über die PPAR-Selektivität, da diese sowohl von hPPARalpha- als auch von hPPARgamma-Agonisten belegt wird. PPARalpha/gamma-Selektivität kann zum einen durch das Besetzen der linken bzw. rechten distalen Bindungstasche erzielt werden, zum anderen durch die Auswahl verschiedener acider Kopfgruppen. Aufgrund unterschiedlicher Aminosäuresequenzen der Bindungstaschen der einzelnen PPAR-Subtypen ergibt sich für die Kopfgruppen der PPAR-Modulatoren im hPPARalpha und hPPARgamma eine durch den sterischen Einfluss von Carboxyl- bzw. TZD-Kopfgruppen verursachtes unterschiedliches Wasserstoffbrückennetzwerk. Im Vergleich mit Pirinixinsäure konnte nur durch die Einführung der größeren Alkylketten (n-Butyloder n-Hexyl) ein stark erhöhter dualer PPARalpha/gamma-Agonismus erreicht werden. Die Substitution des sekundären Amins der Chinolin-6-ylverbindungen durch einen Ether-Sauerstoff in den Substanzen 41 und 42 sowie die Einführung eines Linkers, genauer einer Methylengruppe, zwischen dem Chinolin-6-ylrest und dem Scaffold (Derivate 29 und 36), führt zu einer verminderten Aktivität sowohl an hPPARalpha als auch an hPPARgamma. Eine mögliche Erklärung hierfür könnte sein, dass an entsprechender Position der Liganden-Bindungstasche eine Wasserstoffbrücken-Akzeptor-Position vorhanden ist, welche zur Erhöhung der Affinität beiträgt. Offensichtlich wird, durch die Ausbildung einer Wasserstoffbrücke zwischen der NH-Funktion und einem Wasserstoffbrücken-Akzeptor innerhalb der LBP im Falle von WY 14643 ein hoher Bindungsbeitrag erreicht. Daten bezüglich der Inhibierung der 5-Lipoxygenase demonstrieren, dass 6-Aminochinolinderivate potente 5-LOX-Inhibitoren sind. Die Einführung eines 3,5-Bis-(2,2,2-trifluor-ethoxy)-phenylrestes in 6-Position des alpha-hexylsubstituierten Scaffolds führt zu Ligand 38. Dieser ist ein potenter dualer PPARalpha/gamma-Agonist mit einem EC50-Wert von 11 mikro M an hPPARalpha bzw. 7,7 mikro M an hPPARgamma und ein stark wirksamer 5-LOX-Inhibitor mit einem IC50-Wert von 1 mikro M im 5-LOX-PMNL-Standardassay. Die Substanz 38 wurde in präklinischen Studien eingesetzt. Vier Stunden nach oraler Administration bei Mäusen (n = 6) wurde einen Plasmaspiegel von 40 mikro M erreicht. Im Hinblick auf eine Steigerung der PPAR-agonistischen Aktivität und der Reduktion der hepatotoxischen Eigenschaften der Leitstruktur Pirinixinsäure wurde eine Leitstrukturoptimierung durch Einführung der größeren Alkylketten in alpha-Position zur pharmakophoren Carboxyl-Funktion durchgeführt. Für die inhibierende Wirkung auf die 5-LOX scheint das Chinolin-Strukturelement verantwortlich zu sein, während die Potenz der Inhibition durch das Substitutionsmuster moduliert wird.
Structural biology and life sciences in general, and NMR in particular, have always been associated with advanced computing. The current challenges in the post-genomic era call for virtual research platforms that provide the worldwide research community with both user-friendly tools, platforms for data analysis and exchange, and an underlying e-Infrastructure. WeNMR, a three-year European Commission co-funded project started in November 2010, groups different research teams into a worldwide virtual research community. It builds on the established eNMR e-Infrastructure and its steadily growing virtual organisation, which is currently the second largest VO in the area of life sciences. WeNMR provides an e-Infrastructure platform and Science Gateway for structural biology. It involves researchers from around the world and will build bridges to other areas of structural biology.
Die vorliegende Arbeit befasste sich in erster Linie mit der Regulation des P2X2 Rezeptors (P2X2R) durch Phosphoinositide (PI). P2X Rezeptoren sind durch extrazelluläres ATP aktivierte Kationenkanäle, die ubiquitär unter Vertebraten, v. a. im zentralen wie peripheren Nervensystem exprimiert werden. Bis heute sind 7 verschiedene Untereinheiten dieser Rezeptorfamilie bekannt, die nach homo- oder heterotrimerer Assemblierung unterschiedliche funktionelle Phänotypen ausbilden. Die P2X Rezeptoren sind an einer Vielzahl von physiologischen und pathophysiologischen Prozessen beteiligt. Für ihre Beteiligung am zellulären Signalgeschehen wurde in der Vergangenheit der Begriff der purinergen Signaltransduktion geprägt. PI4,5P2 ist ein zelluläres, an der Innenseite der Plasmamembran verankertes Phospholipid, dem zahlreiche, essentielle Funktionen zukommen. Dass es auch Signalfunktion besitzen kann, wurde erst spät (1980) bekannt; dass es darüber hinaus zudem membranäre Transportsysteme reguliert, konnte erst in den letzten Jahren gezeigt werden. Die ersten Kanäle, für die eine Phosphoniositid (PI)-Beeinflussung nachgewiesen wurde, waren die einwärts gleichrichtenden K+-Kanäle. 2006 wurden die ersten vorläufigen Hinweise publiziert, dass auch die Kanalfunktion der P2X Rezeptoren durch Phosphoinositide beeinflusst werden kann. Darauf aufbauend wurde in der vorliegenden Arbeit der P2X2R nach Expression in Xenopus Oozyten elektrophysiologisch auf eine mögliche Regulation durch PIPns untersucht. Um die in der Oozyte vorliegenden PI-Level gezielt während der Messung ändern zu können, wurde die spannungsgesteuerte Phosphoinositid-Phosphatase Ci-VSP coexprimiert. Ci-VSP, die der PTEN-Phosphatase strukturell sehr ähnlich ist, wurde 2005 aus der Schlauchascidie Ciona intestinalis kloniert. In der veröffentlichten Klonierungsarbeit wurde bereits gezeigt, dass Ci-VSP in der Lage ist, bekannte PI4,5P2-sensitive Membrankanäle, wie z. B. bestimmte K+-Kanäle, spannungsabhängig zu inhibieren. Es konnte in TEVC-Experimenten gezeigt werden, dass die durch Depolarisation induzierte Aktivierung dieser Phosphatase den P2X2 Rezeptorstrom in seiner Desensibilisierung sowohl beschleunigt als auch verstärkt. Dieser Effekt war spannungsabhängig und nahm mit höherer Depolarisation zu. Die ermittelte Spannungsabhängigkeit stimmte dabei mit dem sensitiven Potentialbereich der spannungsgesteuerten Ci-VSP-Domäne, gemessen an ihren gating-Strömen, überein. Der „Ci-VSP-Effekt“ auf den P2X Rezeptor konnte nur in Anwesenheit von ATP, d.h. bei aktiviertem Rezeptor, beobachtet werden. Wurden die Oozyten mit Wortmannin, einem PI4-Kinase(PI4K)-Inhibitor, behandelt, zeigte sich eine vergleichbare Veränderung des Rezeptorstroms. Eine PI4K-Inhibition zielt demnach offensichtlich auf die gleichen Regulationsmechanismen wie die Ci-VSP-Aktivierung. In weiterführenden zellfreien Patch Clamp-Messungen an Oozytenmembranen wurden sowohl Einzelkanal- als auch makroskopische Ströme des P2X2R unter Einfluss verschiedener, intrazellulär verabreichter PIs und PI-beeinflussender Enzyme untersucht. Einzig die Zugabe von PI4,5P2 hatte einen deutlich aktivierenden Einfluss auf den makroskopischen Rezeptorstrom, andere getestete PIs (wie auch PI3-Kinase und PTEN-Phosphatase) zeigten keinerlei Wirkung. Vergleichbare Ergebnisse konnten in vorläufigen Einzelkanal-Messungen an diesem Rezeptor Subtyp beobachtet werden. Da die PI4-Kinase offensichtlich an der beobachteten Beeinflussung der P2X2R Desensibilisierung beteiligt ist, wurde die P2X2-Rezeptorsequenz auf potentielle - über sogenannte SH3-Epitope vermittelte - PI4K-Interaktionsbereiche hin untersucht. Diese SH3-Epitope kommen in vielen zellulären Proteinen vor, um Protein-Protein-Interaktionen zu vermitteln. Nach Sequenzanalyse des maßgeblich am Desensibilisierungsgeschehen beteiligten C-Terminus des P2X2R konnte im distalen Teil ein SH3-Bindungsmotiv lokalisiert werden, das daraufhin durch gerichtete Mutagenese (P2X2-P451A/P454A) unwirksam gemacht wurde. Dieser mutierte Rezeptor verhielt sich in seiner Desensibilisierung wie der Wildtyp nach Wortmannin-Behandlung, zeigte also eine intrinsisch verstärkte Desensibilisierung. Eine Wortmannin-Behandlung der Oozyten, die den mutierten Rezeptor exprimierten, führte hingegen zu keiner weiteren Beeinflussung des Rezeptorstroms. Somit konnte letztlich der Schluss gezogen werden, dass die PI4K, und das mit ihr in direkter Verbindung stehende PI4,5P2, einen maßgeblichen Einfluss auf das Desensibilisierungsverhalten des P2X2R hat. Auf Basis der erarbeiteten Befunde wurde ein kinetisches Reaktionsmodell des P2X2R erstellt, das bisher aufgestellte Modelle mit den Ergebnissen dieser Arbeit vereint, aber auch in teilweisem Gegensatz zu dem von FUJIWARA & KUBO [2006] steht. Des Weiteren wurde im Verlauf dieser Arbeit die reversible Inhibition des P2X2R durch eine Reihe von Aminoglykosid-Antibiotika untersucht. Durch Analyse der Dosis-Wirkungs-Beziehungen, der Spannungs- sowie wie ATP-Konzentrations-Abhängigkeit der Inhibition konnte gezeigt werden, dass es sich dabei um einen nicht-kompetitiven open pore block handelte. Durch weiterführende Untersuchungen an einer nicht-desensibilisierenden P2X2/1 Rezeptorchimäre wurde gezeigt, dass eine Aminoglykosid-Inhibition die ATP-Dissoziation von der Rezeptorbindungsstelle signifikant verlangsamte. Dieser Befund deutet auf eine im Vergleich zum geschlossenen Zustand erhöhte Affinität des offenen Zustands für ATP hin. Neben den hier untersuchten Aminglykosiden sind bislang keine weiteren Substanzklassen bekannt, die den P2X2R durch einen derartigen Mechanismus hemmen.
Redirection of miRNA‐argonaute complexes to specific target sites by synthetic adaptor molecules

Dysregulation of miRNAs is connected with a multitude of diseases for which antagomirs and miRNA replacement are discussed as therapeutic options. Here, we suggest an alternative concept based on the redirection of RISCs to non‐native target sites. Metabolically stable DNA‐LNA mixmers are used to mediate the binding of RISCs to mRNAs without any direct base complementarity to the presented guide RNA strand. Physical redirection of a dye‐labeled miRNA model and of specific miRNA‐programmed RISC fractions present in HeLa extracts is demonstrated by pull‐down experiments with biotinylated capture oligonucleotides.
The title compound, C25H22O5, was obtained by a dehydrogenative carbonylation reaction. It crystallizes with one half-molecule in the asymmetric unit. The molecules have crystallographic C2 symmetry and the two atoms of the carbonyl group are located on the rotation axis. The methoxy groups are coplanar with the benzene ring to which they are attached [C-C-O-C = 1.0 (6)°]. The two furan rings are inclined at 17.3 (3)° with respect to each other and the dihedral angle between the furan ring and the benzene ring is 75.83 (12)°. The crystal structure is stabilized by C-H...O hydrogen bonds. The isolation of transcribed DNA sequences of P815 cells and the partial characterization with respect to their sequence composition and relative rates of enzymatic DNA methylation are reported in this paper. Transcribed regions were purified by affinity chromatography using immobilized heterogenous nuclear RNA of P815 cells. About 10% of total genome was found in this fraction. Reassociation analyses showed differences in sequence composition of transcribed versus non-transcribed DNA fractions. The relative proportion of inverted repeats was doubled in the transcribed fraction whereas ordinary highly repetitive sequences comprising mainly of satellite DNA were found almost exclusively in the non-transcribed regions of the P815 genome. About 70% of transcribed portions corresponds to unique and intermediary DNA sequences. After labelling of cells with L-[Methyl-3H]methionine and [14C]deoxycytidine relative rates of enzymatic DNA methylation were computed for different kinetic components of transcribed and non- transcribed portions of P815 genome. No difference was found except in inverted repeats. In transcribed DNA the relative rate of enzymatic DNA methylation was only about 40% of that of the non-transcribed ones. We have quantitated this hypomethylation and found that there is, in average, about one 5-methylcytosine residue in 100 nucleotides of transcribed inverted repeats, compared to about 2.5 5-methylcytosines in non-transcribed fractions. In view of these data we propose that the enzymatic methylation of inverted DNA repeats negatively controls the transcriptional process in a given genomic region.
A specific class of DNA sequences, the inverted repetitive sequences, forms a double-stranded structure within a single linear polynucleotide chain in denatured DNA. The reassociation process is unimolecular and occurs very fast. Quantitative analyses have shown that these sequences com-E rise about 4-5% of the nuclear DNA of various mammalian cells (P815 mouse mastocytoma, Hela, L cells, Raji and Chang cells, and human embryonic hepatocytes) and are interspersed within sequences of other degrees of repetitiveness.
After labeling the cells with L-[Metnyl-3H]methionine and [14C]deoxycytidine, relative rates of enzymic DNA methylation were computed on the basis of 3H and 14C radioactivities found in py rimidine residues of the nuclear DNA. The results indicate that DNA of inverted repetitive sequences is methylated to a level about 50% higher than the ordinary repetitive sequences and to about 300% higher than the unique and intermediary sequences.
The biological function of the inverted repeats as well as the role of their enzymic hypermethyl ation is unknown.
The sequence complexity of nuclear RNA from mouse liver, mouse spleen and highly malignant P815 mastocytoma was measured by nRNA driven hybridization to unique DNA sequences of P815 cells. The unique DNA sequences represent 63% of the total nuclear DNA of P815 cells and their availibility in hybridization experiments was found to be 76%. Of these sequences 7.8% formed hybrids with nuclear RNA of this cell, about 11.5% with mouse spleen and about 14.5% with mouse liver nuclear RNA. Assuming an asymmetrical transcription, the complexities of these transcripts are 2.8 × 108 nucleotides for mouse P815 mastocytoma, 4.3 × 108 for mouse spleen and about 5.3 × 108 nucleotides for mouse liver.
Cellular specifity of the transcribed information was analyzed in additivity experiments, in which unique DNA sequences, not complementary to the nuclear RNA of one cell were annealed to the nuclear RNAs of the two other tissues/cells. In these experiments most of the nuclear RNA se quences of P815 cells were found to be also present in the nucleus of mouse liver and spleen. Only a small portion of the unique DNA sequences of P815 mastocytoma (about 1.2% corresponding to 4.4 ×107 nucleotides) was found to be complementary only to P815 mastocytoma nuclear RNA.
In den letzten zwei Dekaden wurde die Rolle der RNA in biologischen Prozessen intensiv untersucht. Man erkannte immer deutlicher, dass ihre Funktion über die einfache Vermittlung von Information von der DNA hin zum Protein weit hinausgeht. So spielt sie eine entscheidende Rolle bei der Genexpression und besitzt darüber hinaus auch katalytische Eigenschaften. Die Entdeckung der reversen Transcriptase beim HI-Virus konnte zeigen, dass genetische Informationen nicht nur in Richtung von DNA zu RNA, sondern auch in Entgegengesetzter Richtung übertragen werden kann. Diese Schlüsselrolle in vielen wichtigen biochemischen Prozessen macht die RNA zu einem viel versprechenden Ziel für die Entwicklung neuer Wirkstoffe, um in diese Prozesse eingreifen zu können. RNA bildet eine Vielzahl von stabilen Sekundär- und Tertiärstrukturen aus, die es Proteinen und Antibiotika ermöglicht, sie zu adressieren. Die bis heute wohl mit Ausnahme des Ribosoms und der tRNAs am besten aufgeklärte Struktur einer RNA ist die der HIV TAR-RNA (transaktivierende Region). Ein essentieller Bestandteil für die virale Genexpression ist die so genannte TAR-RNA. Diese befindet sich am 5-Ende aller viraler Transcripte und bildet eine aus 59 Basen bestehende stem-bulge-loop hairpin Struktur. Diese tritt in Wechselwirkung mit dem tat-Protein. Die Grundlage für die Erkennung des hierbei entstehenden Komplexes ist eine Wechselwirkung von tat mit der bulge- und loop-Region von TAR. Durch Interaktion mit der loop- Region kommt es zur Ausbildung eines CyclinT1/tat Komplexes, der im weiteren dafür verantwortlich ist, dass sich die Rate der Transkription um das mehrere hundertfache erhöht. Ziel der vorliegenden Arbeit war es, Liganden zu synthetisieren, die spezifische Bindungen mit der TAR-RNA aus HIV-1 eingehen. Durch eine solche Bindung ist es möglich, den viralen Replikationszyklus zu unterbrechen. Ausgehend von einem relativ rudimentären Design, basierend auf dem Arginin-Fork-Model von Frankel, gelang es, mit Triaminopyrazol und verschiedenen seiner Derivaten dieses Ziel zu erreichen. Nach erfolgreicher Synthese der Zielstrukturen wurden diese in einem FRET-Assay im Hinblick auf ihre Affinität zu der TAR-RNA getestet. 3,4,5-Triaminopyrazol übertraf trotz seiner geringen Größe und Ladung mit einem IC50-Wert von 30 µM die Werte vieler tetrakationischer Tripeptide (FRET). Nach erfolgreicher Bestimmung der TAR-Affinität von Triaminopyrazol wurde seine Wirkung auf HIV-infizierte Hela P4-Zellen untersucht, die ein Tat-TAR-kontrolliertes Reportergen exprimierten. Dabei zeigte Triaminopyrazol eine Inhibierung mit IC50 = 50 µM. Bis zu einer Konzentration von 500 µM traten hierbei keine toxischen Effekte auf. Dies legt die Vermutung nahe, dass es sich bei Triaminopyrazol tatsächlich um einen tat- Antagonisten handelt. Ebenso konnte gezeigt werden, dass die Wirkung von Triaminopyrazol nicht die eines Entryinhibitors ist, sondern dass die Verbindung in der Lage ist, die Zellmembran zu durchdringen.
Pulsed electron-electron double resonance (PELDOR) is a well established method concerning nanometer distance measurements involving two nitroxide spin-labels. In this thesis the applicability of this method to count the number of spins is tested. Furthermore, this work explored the limits, up to which PELDOR data obtained on copper(II)-nitroxide complexes can be quantitatively interpreted. Spin counting provides access to oligomerization studies – monitoring the assembly of homo- or hetero-oligomers from singly labeled compounds. The experimental calibration was performed using model systems, which contain one to four nitroxide radicals. The results show that monomers, dimers, trimers, and tetramers can be distinguished within an error of 5% in the number of spins. Moreover, a detailed analysis of the distance distributions in model complexes revealed that more than one distance can be extracted from complexes bearing several spins, as for example three different distances were resolved in a model tetramer – the other three possible distances being symmetry related. Furthermore, systems exhibiting mixtures of oligomeric states complicate the analysis of the data, because the average number of spin centers contributes nonlinearly to the signal and different relaxation behavior of the oligomers has to be treated explicitly. Experiments solving these problems are proposed in the thesis. Thus, for the first time spin counting has been experimentally calibrated using fully characterized test systems bearing up to four spins. Moreover, the behavior of mixtures was quantitatively interpreted. In addition, it has been shown that several spin-spin distances within a molecule can be extracted from a single dataset. In the second part of the thesis PELDOR experiments on a spin-labeled copper(II)-porphyrin have been quantitatively analyzed. Metal-nitroxide distance measurements are a valuable tool for the triangulation of paramagnetic metal ions. Therefore, X-band PELDOR experiments at different frequencies have been performed. The data exhibits only weak orientation selection, but a fast damping of the oscillation. The experimental data has been interpreted based upon quantitative simulations. The influence of orientation selection, conformational flexibility, spin-density distribution, exchange interaction J, as well as anisotropy and strains of the g-tensor has been examined. An estimate of the spin-density delocalization has been obtained by density functional theory calculations. The dipolar interaction tensor was calculated from the point-charge model, the extension of the point-dipole approximation to several spin bearing centers. Even assuming asymmetric spin distributions induced by an ensemble of asymmetrically distorted porphyrins the effect of delocalization on the PELDOR time trace is weak. The observed damping of dipolar oscillations has been only reproduced by simulations, if a small distribution in J was assumed. It has been shown that the experimental damping of dipolar modulations is not solely due to conformational heterogeneity. In conclusion the quantitative interpretation of PELDOR data is extended to copper-nitroxide- and multi-spin-systems. The influence of the mean distance, of the number of coupled spins, of the conformational flexibility, of spin-density distribution and of the electronic structure of the spin centers has been analyzed using model systems. The insights on model compounds mimicking spin-labeled biomacromolecules – in oligomeric or metal bound states – calibrate the method with respect to the information that can be deduced from the experimental data. The resulting in-depth understanding allows correlating experimental results (from for example biological systems) with models of structure and dynamics. It also opens new fields for PELDOR as for example triangulation of metal centers and oligomerization studies. In general, this thesis has demonstrated that modern pulsed electron paramagnetic resonance techniques in combination with quantitative data analysis can contribute to a detailed insight into molecular structure and dynamics.
Erfolgreich wurde die biochemische Synthese eines Kernbereiches der spleißosomalen U4/U6-RNA etabliert und die Sekundärstruktur mittels NMR-spektroskopischer Methoden charakterisiert. Die Konformationen von zwei molekularen Regeleinheiten, des auf Gramicidin A basierenden Ionenkanals Minigramicidin sowie einem aus zwei cis-Dekalinen aufgebautenmolekularen Schalters konnten in unterschiedlichen Umgebungen mit Hilfe der NMR-Spektroskopie erfolgreich bestimmt werden. Die Synthese des RNA-Konstruktes u4u6a46phh2 erfolgte durch Transkription von plasmidischer Templat-DNA mit T7-Polymerase und anschließender Aufreinigung mittels Gelelektrophorese und Homogenisierung am 3’-Ende mit Hilfe eines passenden Hammerhead-Ribozyms. u4u6a46phh2-RNA kann als Konstrukt für die Synthese von 13C/15N-gelabelter RNA dienen, da die Schneidreaktion und die daraus resultierende RNA definiert ist und die Integrale der Iminoprotonen für eine einzige Konformation der RNA sprechen. Das von Dr. Hans-Dieter Arndt in der Arbeitsgruppe von Prof. Dr. Koert an der Humboldt-Universität zu Berlin hergestellte Minigramicidin ist aus zwei verkürzten Gramicidin A-Einheiten aufgebaut, die in einer Kopf-an-Kopf Anordnung durch einen Bernsteinsäure-Linker kovalent verknüpft sind. In dieser Arbeit wurden die Strukturen von Minigramicidin in zwei unterschiedlichen Umgebungen aufgeklärt: in Benzol/Aceton (10:1, v/v) ohne Zusatz von Kationen und in gesättigter Cäsiumchlorid-Chloroform/Methanol (3:1, v/v)-Lösung. Im ersten Fall findet man ein doppelt helikales linksgängiges Dimer von Minigramicidin mit ca. 5.7 Resten pro Windung. Diese Struktur hat eine Länge von ca. 38 Å und einen Durchmesser von ca. 1.2 Å. Mit Cäsium-Kationen liegt die Verbindung als Monomer vor. Sie bildet eine rechtsgängige pi-Helix mit ca. 6.3 Resten pro Windung. Die Struktur hat eine Länge von 17 Å und einen Durchmesser von ca. 4.5 Å. Es konnte erstmals eine zur ionenkanalaktiven Form des gA ähnliche Konformation eines auf gA basierenden künstlichen Ionenkanals in organischen Lösemitteln nachgewiesen werden. Der von Dr. Michael Karle aus der Arbeitsgruppe von Prof. Koert an der Philipps-Universität Marburg synthetisierte molekulare Schalter besteht aus zwei cis-Dekalineinheiten, die durch einen 14-gliedrigen Bislactam-Ring miteinander gekoppelt sind. Der Schaltprozeß wurde dabei durch die saure Spaltung des Ley-Acetals in 13 ausgelöst. Es wurde für die Verbindung 13 eine all-axial Stellung für den Makrozyklus gefunden. Diese Struktur wird auch durch die gefundenen Kopplungskonstanten gestützt. Nach dem Schaltprozeß wurde die Struktur von 16 ermittelt. Wie erwartet, wurde durch das Lösen der konformativen Klammer ein Doppelringflip im linken Dekalingerüst ausgelöst und durch den Makrozyklus auf das rechte Dekalingerüst übertragen. Die gefundenen ROE-Abstände und Kopplungskonstanten für bestimmte Dekalinprotonen bestätigen die umgeschaltete Struktur.
The following mixed-stack donor/acceptor complexes {D···A}∞ have been crystallized and their structures determined: {hexamethylbenzene···3,5-dicyano-1-nitrobenzene hexamethylbenzene···3,5-dinitro-1-cyanobenzene}∞, {pyrene···3,5-dinitro-1-cyanobenzene}∞, {anthracene···(3,5-dinitro-1-cyanobenzene)2}∞, {N,N-dimethylanilin···3,5-dinitro- 1-cyanobenzene}∞ and { 1-3-phenylenediamine···3,5-dinitro-1-cyanobenzene}∞. Their lattice packing consists of parallel layers, which contain either donors and acceptors as for hexamethylbenzene and pyrene or composite ones as in the 1:2 complex of anthracene with each one of the acceptors above and below its peripheral rings. The isostructural hexamethylbenzene complexes exhibit almost identical packing coefficients as well as a hexagonal coplanar arrangement of the C6(CH3)6 donors. Weak intermolecular van der Waals interactions are also observed between antiparallel cyano substituents. The interplanar n distances range between 334 and 353 pm, i. e. around 340 pm of two van der Waals n radii. In none of the complexes, however, significant structural changes in either the donor or the acceptor components due to the complex formation are observed. In both the crystals as well as in solution, the donor/acceptor complexes exhibit colours between yellow and red; their long-wavelength charge transfer absorption maxima, therefore, correspond to a lowering in excitation energy of only up to 1 eV relative to that of the components. The different charge transfer in the ground and the CT excited states is also discussed referring to other data such as vertical first ionization energies or interplanar distances {D···A}, as well as to results from semiempirical calculations based on the crystal structure data determined and including approximate configuration interaction.
Raney nickel, a highly reactive and air-sensitive solid, if prepared and investigated under oxygen-free conditions, exhibits interesting catalytic properties. Using photoelectron spectroscopy for real-time gas analysis in a flow reactor, the following results are obtained with alkyl and acylhalides: Dehydrohalogenation temperatures are lowered relative to thermal HHal elimination up to 350 K. Monochloro and bromo propanes and butenes yield propene and butadiene, respectively. 1,1-Dichloro ethane or 1,1-dibromo propane only split off one HHal and form chloroethene or 1-bromopropene-2. HCl elimination from 2-methyl propionic acid chloride, expectedly, produces dimethyl ketene. Most interesting, however, is the ring opening of monobromo cyclobutane to 1-bromo-butene-3, observed already at room temperature, which strongly suggests the intermediate formation of a chemisorbed surface carbene at Raney nickel. The formation of hexadiene-1,5 as a by-product in the HCl elimination of 1-chloropropane, i. e. a surface carbene dimer, indicates their presence also in other dehydrohalogenations heterogeneously catalyzed by Raney nickel.

The following mixed-stack donor/acceptor complexes {D · · · A }∞ have been crystallized and their structures determined: { 1 ,2,4,5-tetramethylbenzene · · · tetrabromo-p -benzoquinone}∞ , {hexamethylbenzene · · · tetrabromo-p-benzoquinone}∞ , { ( 1 ,2 ,4,5-tetramethyl-benzene)2 · · · tetrachloro -p -benzoquinone}∞ , {pyrene · · · tetrafluoro-p-benzoquinone}∞ , {pyrene · · · tetrabromo-p-benzoquinone}∞ and {perylene · · · tetrabromo-p-benzoquinone}∞ . They exhibit an interesting lattice packing, especially the 2:1 tripeldecker sandwich of tetrachloro-p-benzoquinone, which crystallizes in a herringbone pattern. Their interplanar distances are around 340 pm, i. e. two van der Waals π radii. None of them , however, exhibits in neither the donor nor the acceptor components significant structural changes due to complex formation. Their colours range from orange-red to black in the crystal and to green in H2CCl2 solution. Their long-wavelengths charge transfer absorption maxim a correspond to a lowering in excitation energy of up to 2 eV relative to that of the components. The different charge transfer in the ground and excited states of the donor/acceptor complexes investigated is further discussed referring to data such as cyclovoltammetric reduction potentials as w ell as to results from semiempirical calculations based on the crystal structure data determined and including configuration interaction.
The neutral title compound, 8,8-bis(dimethylamino)dibenzo-[a,d]-heptafulvene, exhibits a first vertical ionization potential of only 6.98 eV and, therefore, can also be oxidized by AlCl3 in H2CCl2 solution. The radical cation generated shows a complex multiplet signal pattern, which is assigned based on additional ENDOR measurements. The photoelectron (PE) and ESR spectra of the 112 valence electron molecule are interpreted by “pararneter-optimized” HMO and by geometry-optimized MNDO calculations, which both suggest a non-planar π-type ground state with most of the charge and the spin distributed over the dibenzoheptatriene part of the radical cation.
Di(methylthio)acetylene H3CS-C≡C-SCH3 reacts with S2C12 in AlCl3/H2CCl2 solution to the tetra(thiomethyl)thiophene radical cation (H3CS)4C4S·⊕ and with H3CSCl to the tetra(thiomethyl)ethene radical cation (H3CS)2C·=⊕C(SCH3)3. Their ESR spectra are assigned by comparison with literature data or those of analogous products obtained from other acetylene derivatives R-C≡C-R with R = SCH2CH3, CH3, C6H5 as well as based on HMO arguments. The possible course of the oxidative sulfuration is discussed.
The structures of seven di- or tetrasubstituted p-benzoquinone derivatives O=C(XC=CH )2C=O and O=C(XC=CX)2C=O with substituents X = -OCH3, -N(CH2)5, - N(CH2CH2)2O, -Cl, -CN and -⊕N(HC=CH)2C-N(CH3)2 are presented and discussed in comparison with published ones substituted by X = -Si(CH3)3, -C6H5, -N(CH3)2, -⊕N(HC=CH)2CN(CH3)2, -O⊖ , and - NO2. Based on the introduction, in which halfwave-reduction potentials, geometry-optimized quantum-chemical calculations on substituent perturbation and known structural data of p-benzoquinone derivatives are used to characterize their molecular ground states. The structural changes indicate how substituent perturbations might be rationalized. Of the categories defined - imperturbed, donor, donor/acceptor and acceptor perturbed - the donorsubstituted p-benzoquinones do exhibit the largest differences, often called cyanine distorsion. In very satisfactory agreement with extensive semiempirical calculations, all effects determined experimentally are discussed in terms of varying charge distribution. With respect to the biochemical importance of p-benzoquinone derivatives, this first structural summary points out important facets.
From the electron and proton transfer equilibria network of quinones in solution a novel intermediate can be prepared by deprotonation of 2,5-bis(trimethylsilyl)hydroquinone to its monoanion using sodium metal. The sodium salt crystallizes in polymer strings connected via O⊖···(H)O hydrogen bridges, which are capped additionally by twofold dimethoxy-ethanesolvated Na⊕ countercations. The single crystal structure determination reveals one of the shortest O⊕ ··· HO distances observed so far of only 246 pm. MNDO calculations further confirm the assignment of hydroquinone monoanion building blocks in the polymer chain. For structural comparison as well as for attempts of its sodium reduction, 2,5-bis(trimethylsilyl)-p- benzoquinone has been synthesized. Its single crystal structure is reported, which does not show any cyanine distortion.
Chelate complexes of 1,2-dimesitoylbenzene radical anion with alkali metal cations exhibit in aprotic solution extremely large ESR /ENDOR metal coupling constants. For rationalization, structures of both the neutral molecule (H3C)3H2C6 - CO - C6H4 - CO - C6H2(CH3)3, in which the two carbonyl groups are twisted out of the benzene ring plane by dihedral angles of ± 3̄7̄°, and a sodium contact ion quadruple have been determined. One of the dimers [dimesitoylbenzeneH⊖ (Na⊕H2N H2C - CH2NH2)]2, although generated by Na metal mirror reduction of 1,2-dimesitoylbenzene in aprotic DME solution with added ethylendiamine for better electron transfer, surprisingly contains two 245 pm short (!) hydrogen bridges ⊖O ··· (H)O and in addition two solvation bridges e ⊖O ··· Na⊕(H2NH2C - CH2NH2) ··· O⊖. Results of MNDO calculations based on the experimental coordinates support the proposed concept.
Crystals of lemon yellow dipotassium nitranilate and of yellow disodium nitranilate dihydrate have been grown and their structures determined at 290 and 200 K. The six-member- ed, O2N-disubstituted rings show a pronounced cyanine distortion with all four CO bonds identical and the two (OCC(NO2)CO)⊖ chains connected by single CC bonds of each 156 pm length. In the anhydrous K⊕ salt, the ring is planar, but in the Na⊕ hydrate salt it exhibits a twist conformation. Quantum chemical calculations allow to reproduce the structure in every detail, demonstrate strong charge alternation along the cyanine chains with considerable delocalization into the O2N acceptor substituents, and suggest that the rather long connecting CC bonds contain positively charged carbon centers on both ends. In addition, metal ion coordination effects as well as the rather high pKa value of nitranilic acid are rationalized.
1,4-Bis(trimethylsiloxy)benzene has been crystallized both by vacuum sublimation and from «-heptane solution, which each yielded colourless plates with identical monoclinic unit cell dimensions (P2/n, Z = 4). The conformation of C[ symmetry shows the two (H3C)3SiO-substituents to be conrotationally twisted around the O-( C6H4)-O axis by dihedral angles o f ± 60°. According to the photoelectron spectroscopic ionisation pattern and its Koopmans’ assignment, IEVn = -εJAM 1, by AM 1 eigenvalues, the gas phase structure should also be of C, symmetry. The results of geometry-optimized MNDO , AM 1 or PM 3 calculations for the monosubstituted derivative H5C6-OS i(CH3)3 are compared with respect to the quality of their fit to the measured data.
The reduction potentials of 40 aromatic nitro compounds Rπ(NO2)n with Rπ = benzene, naphthalene, anthracene, fluorene and carbazole and n = 1 to 4 nitro groups are determined by cyclic voltammetry in DMF under aprotic conditions. The perturbation by the strongly electron accepting substituents can be rationalized via correlation with HMO eigenvalues. Based on reversibility criteria, the electrochemical behaviour is discussed and the compounds are classified with respect to reversible or irreversible one-electron transfer as well as up to 4 (quasi)-reversible reduction steps. The CV data measured can be used to predict redox reactions of aromatic nitro compounds in inert solvents.
UV/VIS and ESR spectra of electron transfer reaction products in aprotic (cH⊕ < 0,1 ppm) solution can be measured in an especially designed and sealed glass apparatus and provide information on unknown facets of the microscopic pathway through the network of interdependent equilibria. For tetraphenyl-p-benzoquinone in tetrahydrofuran, single-electron reduction by a sodium metal mirror produces a red solution and, unexpectedly, after addition of 2.2.2. cryptand, contact with a potassium metal mirror generates a green (!) one. For both, ESR/ENDOR spectra prove the presence of tetraphenyl-p-benzoquinone radical anion. UV/VIS measurements provide the clue: In the equilibrium revealed by repetetive spectra recording, M·⊖solv + Me⊕solv ⇄ [M·⊖···Me⊖]solv, the radical anion is green (vm = 16900 cm-1) and the contact ion pair red (vm=18900 cm-1 ). On ion pair formation, therefore, the excitation energy of the radical anion increases by 0.25 eV.
In an especially designed and sealed glass apparatus, a combination of UV/VIS and ESR spectroscopy measurements are performed to follow electron transfer reactions in aprotic (cH⊕ < 0,1 ppm) solution. For the sodium metal reductions of the tetracyano-substituted title compounds, the novel technique provides the following detailed information: 1,2,4,5- tetracyanobenzene is uniformly reduced to its radical anion, for which additional geometryoptimized MNDO calculations predict an already significant cyanine disortion. For 7,7,8,8- tetracyano-p-quinodimethane, UV/VIS band shape analysis allows to detect in the saturated THF reduction solution the 16300 cm-1 absorption of the donor/acceptor complex formed in the equilibrium TCNQ·⊖ + TCNQ ⇆ {TCNQ·⊖···TCNQ}, which according to a literature search has been crystallized and structurally characterized in paramagnetic salts such as [Me2⊕ (TCNQ·⊖)2(TCNQ)].
Novel radical anions of trimethylstannyl substituted naphthalenes and their ESR spectra are reported. Both 119 Sn and 117 Sn coupling can be assigned unequivocally. The perturbation of π systems by R3X substituents of group IV b elements X = C, Si, Ge, Sn and Pb is discussed with respect to photoelectron ionization potentials, charge transfer excitations, half-wave reduction potentials and ESR spin distribution.
The photoelectron (PE) spectra of bis(dialkylamino) acetylenes R2N-C≡C-NR2 and of tetrakis(dialkylamino) allenes (R2N)2C=C=C(NR2)2 with R = CH3, C2H5 exhibit characteristic ionization patterns which are assigned to π radical cation states of the two molecular halves twisted against each other. The low first ionization potentials between 7.0 eV and 7.7 eV stimulated attempts to oxidize using AlCl3 in H2CCl2 or D2CCl2. The hyperfine structured ESR spectra observed can be unequivocally assigned to the ethylene radical cations R2N-HC=CH -NR2˙⊕ which are formed from the obviously non-persistent species R2N-C≡C-NR2˙⊕ via a hydrogen transfer. During the oxidation of the dialkylamino-substituted allenes no paramagnetic intermediates could be detected, presumably due to a rapid dimerisation of the allene radical cation (R2N)2C=C=C(NR2)2˙⊕.
The one-electron transfer to large π-delocalized hydrocarbons provides an interesting possibility to crystallize solvent-separated ion-pair salts containing optimally solvated cations. Accordingly, the reduction of 9.9′-bianthryl in aprotic 1.2-dimethoxyethane (DME) solution at a sodium metal mirror allows to grow dark blue, brick-like crystals of its radical anion and threefold DME-solvated sodium cation. The structure of the radical anion is very similar to that recently published for the neutral molecule. According to AM 1 enthalpy hypersurface calculations based on the structural data, the torsion angle between 60° and 120° is determined by the lattice packing and the negative charge is -π-delocalized predominantly within only one anthracene subunit. The counter cation [Na⊕(DME)3], reported only three times so far, shows a sixfold propeller-like coordination of approximate D3 skeletal symmetry with contact distances Na⊕···O between 232 and 243 pm and angles ≮ONa⊕O varying between 69° and 159°. Due to the small repulsion between the chelating DME molecules, the isodesmically calculated Na⊕ solvation enthalpy is more negative than that of the analogous tetrahydrofuran complex [Na⊕(THF)6] - as confirmed by the laboratory experience that salts of less stable anions are preferentially crystallized from a strongly cation solvating DME solution.
Tetraphenyl-p-benzoquinone, according to its single crystal structure, shows some steric congestion: its quinone ring is distorted by 7° to a chair conformation, and its phenyl substituents are twisted around their CC axes between 46° and 72°. The half-wave reduction potentials of -0.57 and -1.25 V in acetonitrile confirm negligible π interaction of the phenyl substituents. Addition of alkalimetal tetraphenylborate salts lowers the second reduction potential due to contact ion formation, which can be confirmed by UV/VIS spectra recorded under aprotic conditions. Extensive ESR/ENDOR investigations prove the formation of the following species in THF solution: Tetraphenyl-p-benzosemiquinone radical anion contact ion pairs [M·⊖ Me⊕solv]' (Me⊕: Li⊕, Na⊕, Rb⊕, Cs⊕) and contact triple ion radical cations both with identical cations [M·⊖ (Me⊕solv)2]·⊕ (Me⊕: Li⊕, Na⊕, Cs⊕) and different cations [M·⊖ (Li⊕solv)(Me⊕solv)]·⊕ (Me⊕: Na⊕, Cs⊕). Addition of crown ethers can lead to external solvation of the Me⊕ counter cations, whereas cryptands form internal solvation complexes. The radical anion of 2,6-diphenyl-p-benzosemiquinone adds cations at its phenyl-free molecular half. The radical anion salt [tetraphenyl-p-benzosemiquinone·⊖ (Na⊕(tetrahydropyrane) 2)] could be crystallized and its structure determined at 200 K. In agreement with the Hirota sign rules for contact radicals in solution, the Na⊕ ion is found 62 pm above the π plane and 29° outside the axis of the CO bound, which is elongated due to one-electron reduction by 5 pm to 127 pm.
Cyclovoltammetric measurements of solutions containing the rather basic tetra-(2′-pyridyl)pyrazine allow to detect even traces of water and thus can be used as a touchstone for aprotic (cH⊕ < 1 ppm) conditions. On exchange of the “innocent” tetrabutylammonium R4N⊕ as supporting electrolyte cation by “interactive” ones such as Li⊕) or Na⊕, considerable changes in the reduction potentials are observed due to ion pair formation.
The structurally different radical anions M⊖ of peralkylated 1-sila-2,5-diazacyclopentane-3,4-dithione and of tetrakis(isopropylthio)-p-benzoquinone are generated by reduction with potassium/2.2.2-cryptand under aprotic conditions in THF solution. On addition of Li⊕B(C6H5)4⊖, both form hitherto elusive sulfur-containing contact ion pairs, which are characterized by their ESR/ENDOR spectra.
Reduction of naturally occurring para-and ortho-benzoquinone derivatives M to their respective radical anions M·⊖ can be accomplished under largely aprotic conditions either by cautious low-temperature reaction in THF containing an excess of (2.2.2) cryptand at a potassium mirror or by using the "mild" single electron transfer reagent tetrabutylammonium boranate R4N⊕BH4⊖ in DMF. On addition of soluble alkali tetraphenylborates Me⊕[B(C6H5)4]⊖ , their hitherto unknown radical ion pairs [M·⊖ Me⊕]· and/or triple ion radical cations [Me⊕M·⊖Me⊕]·⊕ form, which might be of biological relevance in molecular carrier and "turn off -turn on" switch processes. On addition of metal perchlorates Me⊕n(ClO4⊖)n with multiply charged counter cations Me⊕n the respective paramagnetic species [M·⊖Me⊕n]·(n-1)⊕ result. Assuming exclusive one-electron transfer reductions without any redox fragmentation reactions, ESR, ENDOR and GENERAL TRIPLE spectra are presented and discussed for the following radical anions and radical ion pairs: mitomycin C (M·⊖ and [M·⊖Mex⊕]·(x-1)⊕ with Me⊕ = Li⊕, Na⊕), streptonigrine (M·⊖ and [M·⊖Lix⊕]·(x-1)⊕), Entobex® (M·⊖ and [M·⊖Me⊕n]·(n-1)⊕ with Me⊕n = Li⊕, Na⊕, Cd⊕⊕, (H5C6)2Tl⊕) as well as brucinequinone ([M·⊖ Me⊕n]·(n-1)⊕ with Me⊕n = Li⊕, Cd⊕⊕, Pb⊕⊕, La⊕⊕⊕).
The radical anion of dimesityltetraketone (ERed, I = -0.40 V) is easily generated in THF by potassium mirror/[2.2.2]-cryptand reduction. Its contact ion pairs with Na⊕, Cs⊕ and Ba⊕⊕ counter cations, prepared in THF solution by single electron transfer from the respective metals, are characterized by their ESR/ENDOR spectra, which exhibit temperature-dependent metal couplings of aNa⊕ = 0.061 mT (190 K), aCs⊕ = 0.021 mT (190 K), and aBa⊕⊕ = 0.145 mT (295 K).
Ion pairs of 1,10-phenanthrolin-5,6-dione radical anion [M · ⊖Me⊕n] ·⊕(n−1) with Me⊕n = Mg⊕⊕, Ca⊕⊕, Sr⊕⊕, Zn⊕⊕, Cd⊕⊕, Pb⊕⊕ and La⊕⊕⊕ are advantageously prepared in aprotic DMF solution containing appropriate metal salts Me⊕nX⊖ by using the ‘mild’ single-electron reducing agent tetra(n-butyl)ammonium-boranate R4N⊕BH4⊖ . For comparison, the ‘naked’ radical anion with the largely interaction-free [K⊕(2.2.2)-cryptand]⊕ counter cation is chosen, which is formed on reduction with potassium in THF solution of (2.2.2)-cryptand. Addition of excess Na⊕[B(C6H5)4]⊖ to the reduction solution only yields a solvent-separated ion pair (M · ⊖)DMF ··· (Na⊕)DMF, whereas in the presence of multiply charged counter cations Me⊕n the respective contact ion pair radical cations [M · ⊖Me⊕n] · ⊕(n−1) are formed. Their g values decrease with increasing nuclear charge of Me⊕n and their metal-s-spin densities increase with the effective counter cation charge n⊕/rMe⊕n. The ESR /ENDOR data recorded suggest Me⊕n complexation by the δ⊖OC -COδ⊖ chelate tongs and the ion pair stability, which is modified by the dielectric properties of the solvent used, may be rationalized by the Coulombic attraction between the radical anion M · ⊖ and the counter cations Me⊕n.
Conditions for ENDOR measurem ents of organosulfur radical cations are discussed and tested. The one electron oxidation of a variety of aromatic sulfur com pounds comprising benzene-1,2-dithiole, 1,4-dithiine, thianthrene and diphenylsulfide derivatives as well as 33S isotope-marked bis(2,5-dimethoxyphenyl)disulfide is accomplished using the oxygen-free, powerful and selective AlCl3/H2CCl2 reagent. Partly with substantial structural changes, paramagnetic M⊕ species of 1,2-benzodithiete, 1,4-dithiine, thianthrene and diphenyl sulfide result. Their temperature-dependent ENDOR signal patterns provide numerous information e.g. on radical cation structure and dynamics, on the rather high sulfur spin populations or on the spin rotation interaction dominated relaxation behaviour. Accordingly, to obtain optimum ENDOR effects in organosulfur radical cations low temperature measurements are required, and especially for still undiscovered 33S ENDOR couplings, small g factor anisotropies and 33S spin densities appear to be necessary.
For the first time, 107,109Ag ENDOR measurements in solution are reported. In addition, the formation of the known paramagnetic contact ion pair [Ag⊕(PR3)2(R2H2C6O2·⊖] on reduction of 3,5-di(tert-butyl)-o-benzoquinone in THF solution containing soluble silver salts and triphenylphosphine is studied by cyclic voltammetry.
The sodium salt of the most simple polynitro-substituted hydrocarbon anion. Na⊕⊖C(NO2)3, (for a hazard warning cf. [***]) crystallizes from ether solutions without and with addition of 18-crown-6 either in a polymer band. [(Na⊕⊖C(NO2)3)dioxane]∞, or as a solvent- separated ion pair, [(Na⊕/18-crown-6)(THF2]⊕[(Na⊕/18-crown-6)(O2N-C⊖(NO2)2)2]⊖. The Na⊕ cations are each 8-fold coordinated in hexagonal bipyramidal arrangement. According to extensive quantum-chemical calculations based on the structure coordinates, the formation of these novel salts can be traced back to the charge distribution in the anions ⊖C(NO2)3. which due to negatively charged oxygen centers are favorable complex ligands. The structure determining effects of solvation are discussed.
The reversible one-electron insertion into mono- and 1,4-di-substituted benzene derivatives is favored by dialkoxyboron and especially by dialkylboron groups. The assumption that it should be the symmetric e2u benzene molecular orbital which is occupied in the resulting radical anions can be supported by comparison of ESR coupling constants.
The PE spectra of the nitrogen-rich title compounds cyanogen azide NC-N3, azodicarbonitrile NC - N = N - CN, azidoacetonitrile NC - H2C - N3, tetrazolo[1,5-a]pyridine (H4C5N)(N )3 and trimethylenetetrazole (H2C)3(CN4) are presented and assigned by radical cation state comparison with related compounds or by Koopmans’ correlation with MNDO eigenvalues. In a low pressure flow system the compounds decompose at higher temperatures, with elimination of the thermodynamically favorable N2 molecule. PE-spectroscopic real-time analysis reveals as further products: NC - N3 → C∞, NC - N = N - CN → NC - CN , NC - H2C - N3 → 2HCN (+ traces NC - HC = NH?) and (H2C)3(CN4) → H2C = N - CN + H2C = CH2. For tetrazolo[1,5-a]pyridine, a preceding ring opening to the corresponding 2-azidopyridine is observed.
Trifluoromethyl azide decomposes in a low-pressure flow system at rather high temperatures by splitting off N2. The nature of the resulting products depends largely on the wall material of the pyrolysis tube: using molybdenum above 1120 K, FCN is observed exclusively. Neither F2C=NF nor F3C-N=N-CF3 can be detected as intermediates by comparing their PE spectra with those continuously recorded while increasing the temperature. F3C-N = N - CF3 fragments already at 870 K to give N2 and F3C-CF3. The PE spectra of F3CN3 and F2C=NF are assigned based on MNDO calculations.

The photoelectron spectra of tricyanomethane derivatives X-C(CN)3 with substituents X = H, CH3, Br and C6H5 have been recorded and are assigned based on MNDO calculations as well as on radical cation state comparison with the iso(valence)electronic P(CN)3, within the series of cyanomethanes H4-nC(CN)n, and with each other. For HC(CN)3, no traces of the isomeric dicyano, ketimine HN = C=C(CN)2 are detected in the gas phase. Tricyanomethylbenzene, H5C6-C(CN)3, exhibiting the highest first ionization energy of any known singly acceptor substituted phenyl derivative, demonstrates the tremendous electron withdrawing effect of the -C(CN)3 group.
The HCl elimination from β-chloroethyl azide (1-azido-2-chloroethane) over potassium tert. butanolate at 350 K in a low pressure flow system is optimized using PE spectroscopic real-time gas analysis. The highly explosive vinyl azide formed can be purified by cool-trapping the by-products. Its subsequent and virtually hazard-free pyrolysis yields 2H-azirine, which can be isolated at temperatures below 240 K.
In contrast, the direct pyrolysis of β-chloroethyl azide requires temperatures above 710 K and results in a simultaneous split-off of both HCl and N2, yielding acetonitrile as the main thermolysis product. No intermediates such as β-chloroethanimine or ketenimine are observed, a result which is interpreted in terms of chemical activation.
Organodisulfide radical cations R2S2′⊕ and R2C2S2 ′⊕ can be generated from aliphatic as well as aromatic cyclic polysulfides in AlCl3/H2CCl2 solutions and characterized by their ESR spectra. Examples presented are the oxidations of 1,2,3-trithiolanes to 1.2-dithiolane radical cations, in which energetically favored planarized 3 electron/2 center bonds are formed.
Thermal decompositions of azo compounds in the gas phase under reduced pressure are further investigated using photoelectron spectroscopic gas analysis. Passing diallyl, diphenyl and phenylmethyl derivatives either through a short-pathway pyrolysis (SPP) apparatus or through an external thermal reactor (ETR) results in the following fragmentations: Under nearly unimolecular conditions (SPP, 10-4 mbar pressure), diallyldiazene decomposes above 600 K to N2 and hexadiene-1,5 with the allyl radical as a detectable intermediate. The PE spectra recorded for diphenyldiazene above 1000 K (ETR, 1-2 mbar pressure) show N2, benzene, as well as traces of diphenyl. Phenylmethyldiazene yields above 800 K (SPP) predominantly N2, toluene, diphenyl and ethane with the methyl radical as the only detectable intermediate. Insertion of quartz wool into the pyrolysis tube (ETR) lowers the fragmentation temperatures, and in addition, above 850 K, HCN and aniline are PE spectroscopically identified. Surprisingly, this second reaction channel can be heterogeneously catalyzed: phenylmethyldiazene decomposes under 10-2 mbar pressure at a [Ni/SiO2] catalyst surface selectively to HCN and aniline.
In the pyrolysis of 1,2,3-benzoselenodiazole using a short-distance furnace, a short-lived intermediate is detected photoelectron spectroscopically. Mass spectra recorded under similar conditions suggest an isomer C6H4Se rearranging to the more stable final product 6-fulveneselone. The ionization pattern obtained by computerized spectra stripping is assigned to benzselenirene by molecular radical cation state comparison based on MNDO calculations.
Tetraphenylbutatriene is reduced under aprotic conditions to its ESR/ENDOR-spectroscopically characterized radical anion and to its dianion, with both electron transfers quasireversible according to cyclovoltammetric measurements. The alkali cation salts, the red contact ion pair [(H5C6)4C4·⊖][Na⊕ (H3COCH2CH2OCH3)3] and the dark violet contact ion triple [(H5C6)4C4⊖⊖][Li⊕(H3COCH2CH2OCH3)3]2 can be prepared by single electron reduction at a sodium metal mirror or by twofold de-protonation of 1,1,4,4-tetraphenylbutyne-2 using lithium-n-butyl. Their single crystal structures as well as that of the parent acetylene have been determined at low temperatures. The essential structural changes observed are the twisting of both molecular halves (H5C6)2CC relative to each other with increasing negative charge. The simultaneously resulting bond alternancy >C = C = C = C< → >C⊖ - C ≡ C⊖ - C < within the cumulene chain is discussed based on MNDO calculations for the structures determined.
The two-electron reduction of tetraphenyl-p-quinodimethane M via its radical anion M⊖ to its dianion M⊖⊖ is explored both by cyclovoltammetry and ESR/ENDOR spectroscopy. Contact of the diglyme solution with added 15-crown-5 under aprotic conditions with a sodium metal mirror yields black crystals of a solvent-separated contact ion triple [M⊖⊖][Na⊕(OCH2CH2)5(H3CO(CH2CH2O)2CH3)]2. The two-electron-insertion into the pquinodimethane derivative R2C⊖=C(HC=CH)2C=CR2 changes its structure drastically to that of a twofold carbanion substituted benzene, R2C⊖ -(C6H4)- ⊖CR2. MNDO calculations provide a rationale for both the tremendous solvation of a Na⊕ center coordinated to seven oxygen centers of 15-crown-5 and of one diglyme molecule and the structural changes as well as the charge distribution in the unique Tetraphenyl-p-quinodimethane dianion (H5C6)2C⊖-(C6H4)- ⊖C(C6H5)2, in which the two negative charges are largely localized at the carbanion center of the benzene -substituents.
The lysosomal polypeptide transporter TAPL belongs to the superfamily of ATP-binding cassette transporters. TAPL forms a homodimeric transport complex, which translocates oligo- and polypeptides into the lumen of lysosomes driven by ATP hydrolysis. Although the structure and the function of ABC transporters were intensively studied in the past, details about the single steps of the transport cycle are still elusive. Therefore, we analyzed the coupling of peptide binding, transport and ATP hydrolysis for different substrate sizes. Although longer and shorter peptides bind with the same affinity and are transported with identical Km values, they differ significantly in their transport rates. This difference can be attributed to a higher activation energy for the longer peptide. TAPL shows a basal ATPase activity, which is inhibited in the presence of longer peptides. Uncoupling between ATP hydrolysis and peptide transport increases with peptide length. Remarkably, also the type of nucleotide determines the uncoupling. While GTP is hydrolyzed as good as ATP, peptide transport is significantly reduced. In conclusion, TAPL does not differentiate between transport substrates in the binding process but during the following steps in the transport cycle, whereas, on the other hand, not only the coupling efficiency but also the activation energy varies depending on the size of peptide substrate.
The ATP-binding cassette transporter TAPL translocates polypeptides from the cytosol into the lysosomal lumen. TAPL can be divided into two functional units: coreTAPL, active in ATP-dependent peptide translocation, and the N-terminal membrane spanning domain, TMD0, responsible for cellular localization and interaction with the lysosomal associated membrane proteins LAMP-1 and LAMP-2. Although the structure and function of ABC transporters were intensively analyzed in the past, the knowledge about accessory membrane embedded domains is limited. Therefore, we expressed the TMD0 of TAPL via a cell-free expression system and confirmed its correct folding by NMR and interaction studies. In cell as well as cell-free expressed TMD0 forms oligomers, which were assigned as dimers by PELDOR spectroscopy and static light scattering. By NMR spectroscopy of uniformly and selectively isotope labeled TMD0 we performed a complete backbone and partial side chain assignment. Accordingly, TMD0 has a four transmembrane helix topology with a short helical segment in a lysosomal loop. The topology of TMD0 was confirmed by paramagnetic relaxation enhancement with paramagnetic stearic acid as well as by nuclear Overhauser effects with c6-DHPC and cross-peaks with water.
Eine in vivo Modifizierung von Blutstammzellen wäre für eine Reihe gentherapeutischer Therapieansätze vorteilhaft. Dies würde voraussetzen, dass retrovirale Vektoren gezielt auf Blutstammzellen ausgerichtet werden können. Für dieses sogenannte Zelltargeting bietet sich das vom Milznekrose-Virus von Vögeln (SNV) abgeleitete Vektorsystem an, bei dem die Rezeptorbindungsdomäne des Env-Proteins modifiziert werden kann. Im Rahmen der vorliegenden Arbeit sollte ein SNV-basierter retroviraler Zelltargeting-Vektor entwickelt werden, der einen selektiven Gentransfer in die primären humanen CD34-positiven hämatopoetischen Zellen ermöglicht. Zur weitergehenden Charakterisierung des SNV-Vektorsystems sollte geklärt werden, ob das Env-Protein des SNV ein mit anderen gamma-retroviralen Env-Proteinen vergleichbares R-Peptid aufweist, dessen mögliche Rolle bei viralem Zelleintritt ebenfalls untersucht werden sollte. Um eine Zielzell-Spezifität des SNV-Vektors zu erreichen, wurde die gesamte SU-Domäne des SNV-Env-Proteins mit einem einkettigen Antikörperfragment (scFv) ersetzt, das gegen das CD34 Molekül gerichtet ist,. Mit diesem modifizierten Env gelang es, [(antiCD34-TM)SNV]-Vektorpartikel herzustellen, die spezifisch CD34-positive Zellen transduzierten. Essentiell für die Erzeugung solcher Vektoren war die Etablierung einer stabilen Verpackungszelllinie, die Vektorpartikel mit einem Titer von 2x105 i.E./ml produzierte. In Transduktionsexperimenten mit verschiedenen Zelllinien wurde gezeigt, dass [(antiCD34-TM)SNV]-Vektoren eine deutliche Präferenz für CD34+-Zellen und nicht für CD34--Zellen besitzen, wobei der Unterschied in der Transduktionseffizienz zwischen CD34-positiven und –negativen Zellen um den Faktor 100 lag. [(antiCD34-TM)SNV]-Vektoren waren in der Lage, den Reportergentransfer auch in primäre humane Stammzellen zu bewirken. Hierzu wurde ein Transduktionsprotokoll so optimiert, dass die aus dem Nabelschnurblut isolierten CD34+-Zellen transduziert werden konnten. Der für diese Zielzellen bestimmte Vektortiter betrug bis zu 2x106 i.E./ml. In einem Gemisch von primären CD34+- und CD34--Zellen konnte der Vektor zwischen dem Target- und Nontarget-Zellen unterscheiden. Somit wurde zum ersten Mal nicht nur Spezifität, sondern auch Selektivität des SNV-Vektorsystems demonstriert. Dieses Ergebnis ist für eine Weiterentwicklung des Vektors für die in vivo Anwendung in Rahmen einer Gentherapie eine wichtige Voraussetzungen. Im zweiten Teil der Arbeit wurde der Fusionsvorgang bei Virus-Eintritt näher untersucht. Anlass dafür war die experimentelle Beobachtung, dass das Env-Protein des SNV bei der Virusknospung von einer viralen Protease innerhalb der zytoplasmatischen Domäne proteolytisch gespalten wird. Ein Sequenz-Vergleich des SNV TM-Proteins mit dem MLV TM-Protein ergab Hinweise darauf, dass es sich um die Abspaltung des sogenannten R-Peptides analog zu MLV handeln könnte. Die Expression von SNV-Env- Mutanten mit einem entsprechend verkürzten C-Tail (Env delta R) führte zur Synzytien-Bildung. Die bildung hochfusogener Oberflächenhüllproteine durch die Abspaltung des R-Peptids konnte auch für andere gamma-Retroviren gezeigt werden. Die Synzytienbildung konnte quantitativ unter den Env delta R-Varianten verschiedener gamma-Retroviren in einem etablierten Fusionsassay verglichen werden. Das Env delta R des endogenen Retrovirus des Schweins (PERV) des Typs A erwies sich als potentestes Fusionsagens. Als Folge der Ergebnisse der vorliegenden Arbeit wurde postuliert, dass die Abspaltung des R-Peptides ein allgemeiner Mechanismus bei der Partikelreifung der gamma-Retroviren ist und eine fusionsregulierende Rolle besitzt. Eine Weiterentwicklung fusionsaktiver Env-Varianten als mögliche therapeutische Gene für eine Tumor-Gentherapie ist somit diskutierbar.
Das wachsende Verständnis für das fein abgestimmte Zusammenspiel aus Struktur und Funktion von Nukleinsäuren resultiert aus unzähligen Forschungsprojekten. Forschende stehen dabei vor der Herausforderung, dass die zu untersuchenden Oligonukleotide sowohl modifiziert als auch in ausreichender Menge und Reinheit dargestellt werden müssen. Die chemische Festphasensynthese ist ein bewährtes Mittel zur Synthese hochmodifizierter DNA und RNA. Allerdings werden Oligonukleotide mit zunehmender Länge unzugänglicher, da die einzelnen Kupplungsreaktionen nicht quantitativ ablaufen, was zu schwer abtrennbaren Abbruchsequenzen führt. Hinzu kommt, dass während der chemischen Synthese harsche Reaktionsbedingungen nötig sind, denen die gewünschten Modifikationen standhalten müssen. (Chemo-) enzymatische Methoden können diese Hürden überwinden und somit den Zugang zu biologisch interessanten, längeren modifizierten Sequenzen ermöglichen. Jedoch erfolgt der enzymatische Einbau von Modifikationen ohne aufwendige Optimierung lediglich statistisch verteilt. Um weitere Erfolge im Bereich der Strukturaufklärung zu erzielen, werden somit Synthesemethoden benötigt, die sich zum positionsspezifischen Einbau von Modifikationen eignen und gleichzeitig den Zugang zu längeren Oligonukleotiden ermöglichen. Zur Untersuchung der Zusammenhänge zwischen Struktur und Funktion haben sich in den letzten Jahren lichtadressierbare Verbindungen als gefragte Modifikationen erwiesen. Der Einsatz von Licht als mildes, nicht-invasives Auslösesignal stellt besonders im biologischen Kontext eine interessante Herangehensweise dar. Um hochwertige Aussagen über das Verhalten von Oligonukleotiden in komplexer biologischer Umgebung machen zu können, muss durch die gezielte Platzierung lichtaktivierbarer Verbindungen ein effizientes AN/AUS-Verhältnis geschaffen werden. Der Einbau photolabiler Schutzgruppen erlaubt eine vorübergehende Beeinflussung der Oligonukleotidstruktur, die durch Abspaltung der Schutzgruppe irreversibel (re-) aktiviert werden kann. Im Gegensatz dazu ermöglicht der Einbau von Photoschaltern eine reversible Adressierbarkeit durch Isomerisierungsprozesse. Die Synthese komplexer gezielt-markierter Oligonukleotide erfolgt zumeist chemisch und ist daher längenlimitiert.
Ziel dieser Doktorarbeit war es, beide Fragestellungen zu vereinen und eine chemo-enzymatische Methode zur RNA-Synthese zu untersuchen, die zum einen die positionsspezifische Modifizierung mit lichtaktivierbaren Einheiten erlaubt und darüber hinaus die Längenlimitierung der chemischen Festphasensynthese überkommt. Im Zentrum der Methode stehen drei enzymatische Reaktionsschritte zum Einbau von photolabil- und photoschaltbar-modifizierten Nukleosid-3‘,5‘-Bisphosphaten: I) eine 3‘-Verlängerung, in der die modifizierten Bisphosphate mit T4 RNA Ligase 1 mit dem 3‘-Ende einer RNA verknüpft werden; II) die Dephosphorylierung des 3‘-Phosphats mit Shrimp Alkaline Phophatase und III) die Verknüpfung der 3‘-terminal modifizierten RNA mit einem zweiten 5‘-phosphorylierten RNA-Fragment, wodurch eine Gesamtsequenz mit gezielt platzierter Modifikation entsteht (Abb. I).
Im ersten Teilprojekt wurden in kollaborativer Arbeit zunächst benötigte photolabile NPE- und photoschaltbare Azobenzol-C-Nukleosid-3‘,5‘-Bisphosphate synthetisiert und grundlegende Bedingungen der enzymatischen Reaktionen erarbeitet. Hierbei konnte der enzymatische Syntheseansatz erfolgreich in Lösung umgesetzt und der chemo-enzymatische Einbau aller synthetisierten Bausteine nachgewiesen werden. Aufbauend auf diesen Erkenntnissen wurde die Methode in eigenständigen Arbeiten weiterverfolgt, um den multiplen Einbau NPE-modifizierter Nukleosid-3‘,5‘-Bisphosphate in direkter Nachbarschaft sowie deren Einbau in DNA/RNA-Mixmere mit Phosphodiester- oder Phosphorthioatrückgrat zu untersuchen. Es konnte gezeigt werden, dass die verwendeten Enzyme neben lichtaktivierbaren Modifikationen zusätzliche Anpassungen der Phosphateinheit sowie unterschiedliche Ribosebausteine in Kombination tolerieren. Da exogene RNA schnell von Exonukleasen abgebaut und somit unwirksam wird, werden zahlreiche stabilisierende Anpassungen an synthetischen RNAs vorgenommen. Zu den häufigsten zählen Phosphorthioate und Modifikationen der Ribose. Mit der erfolgreichen Modifikation der chimären Oligonukleotide eröffnet die erarbeitete Methode einen wichtigen Zugang zu therapeutisch interessanten Oligonukleotiden. Ein weiterer wichtiger Schritt in Richtung biologisch relevanter Anwendungsmöglichkeiten konnte mit der Synthese, Charakterisierung und Umsetzung eines DEACM-geschützten Uridin-3‘,5‘-Bisphosphates (pUDEACMp) errungen werden. Im Vergleich zur verwendeten NPE-Schutzgruppe ist das Absorptionsspektrum der DEACM-Schutzgruppe bathochrom-verschoben, was eine Abspaltung mit Wellenlängen > 400 nm erlaubt. Dadurch können Zellschäden vermieden und Oligonukleotide mit NPE- und DEACM-Modifikation wellenlängenselektiv angesprochen werden...
By means of differential thermoanalysis, the miscibility of the main polar tetraether lipid of Thermoplasma acidophilum with two ester lipids, dipalmitoyl phosphatidylcholine and dipalmitoyl phosphatidylglycerol, resp., in the presence of excess water was studied. It is shown that with increasing fraction of tetraether lipid in the mixture, the transition range of dipalmitoyl phosphatidylcholine is broadened and the temperature of the maximum heat flow (Tm) is shifted to lower temperatures; furthermore, the enthaply change (ΔH) of the transition declines. Similar results were obtained with mixtures of tetraether lipid with dipalmitoyl phosphatidylglycerol. It is therefore concluded that the main polar tetraether lipid of Thermoplasma acidophilum , which essentially forms monomolecular layers, is able to form stable common phases with bilayer-forming ester lipids. Miscibility of the tetraether lipid with dipalmitoyl phosphatidylglycerol, which are both monovalent anions at neutral pH, is also observed in the presence of high proton or calcium ion concentrations.
Mitochondrial ATP synthases form dimers, which assemble into long ribbons at the rims of the inner membrane cristae. We reconstituted detergent-purified mitochondrial ATP synthase dimers from the green algae Polytomella sp. and the yeast Yarrowia lipolytica into liposomes and examined them by electron cryotomography. Tomographic volumes revealed that ATP synthase dimers from both species self-assemble into rows and bend the lipid bilayer locally. The dimer rows and the induced degree of membrane curvature closely resemble those in the inner membrane cristae. Monomers of mitochondrial ATP synthase reconstituted into liposomes do not bend membrane visibly and do not form rows. No specific lipids or proteins other than ATP synthase dimers are required for row formation and membrane remodelling. Long rows of ATP synthase dimers are a conserved feature of mitochondrial inner membranes. They are required for cristae formation and a main factor in mitochondrial morphogenesis.
Im Rahmen dieser Arbeit wurden zum Vergleich die Strukturen der ATP-Synthasen von Arabidopsis thaliana, Asparagus officinalis, Allium cepa, Helianthus annus, Solanum tuberosum, Bos taurus und Saccharomyces cerevisiae gelöst. Die ATP-Synthase von S. cerevisiae konnte mit einer Auflösung von 19 Å gelöst werden. Der Winkel zwischen den zwei ATP-Synthase-Monomeren in dem ATP-Synthase-Dimer hatte für jede Spezies einen bestimmten Wert. Dieser Winkel änderte sich innerhalb einer Spezies nur wenig im Gegensatz zu Untersuchungen mit Einzelpartikelanalyse.
Die ATP-Synthase-Dimere aus den untersuchten Spezies besitzen unterschiedliche Winkel zwischen 78˚ und 122˚. Der Winkel des ATP-Synthase-Dimers aus S. tuberosum (122˚) viel größer als der in anderen Pflanzen (~98˚), B. taurus (105˚) und S. cerevisiae (78˚). Die Proben von S. tuberosum und B. taurus waren jedoch dünner, was den Winkel eventuell beeinflussen könnte. Um dies auszuschließen müssen in Zukunft weitere Untersuchungen durchgeführt werden.
Des Weiteren wurde im peripheren Stiel der ATP-Synthasen von allen Pflanzenspezies eine Dichte entdeckt, die in B. taurus und S. cerevisiae nicht vorhanden ist. Die Dichte könnte durch eine zusätzliche Untereinheit oder veränderte Untereinheit im Vergleich zu B. taurus und S. cerevisiae kommen.
Weiterhin wurde die Bildung von Reihen aus ATP-Synthase-Dimeren untersucht. Es wurden ATP-Synthase-Dimere von Polytomella sp. gereinigt und in Lipid rekonstituiert. Es wurde das ATP-Synthase-Dimer von Polytomella sp. verwendet, da dieses besonders stabil ist und während der Reinigung nicht zum ATP-Synthase-Monomer zerfällt. Zur Rekonstitution wurde die milde GRecon-Methode verwendet. Hierbei werden Membranproteine in einem Zuckergradienten gleichzeitig in Lipid rekonstituiert und nach ihrer Dichte getrennt. Abhängig von der Dichte der Proteoliposomen ist die Konzentration an Membranproteinen unterschiedlich. In Proteoliposomen mit einer hohen Konzentration bilden sich dünne Schichten in denen die ATP-Synthase-Dimeren Zickzack-Muster formen. Dies deutet darauf hin, dass das ATP-Synthase-Dimer die Membran verformt. In Proteoliposomen mit einer niedrigeren Konzentration an ATP-Synthase-Dimeren wurden runde Vesikel detektiert, in denen die ATP-Synthase-Dimere lange Reihen bilden und die Membran innerhalb jedes ATP-Synthase-Dimer ebenfalls verformt ist. Molekulare Simulationen bestätigen dieses Ergebnis.
Zudem wurde das ATP-Synthase-Dimer in zwei verschiedene Lipide ohne Cardiolipin rekonstituiert, da Cardiolipin ein Lipid ist welches in der bakteriellen und mitochondrialen Membran gefunden wurde und in hohen Konzentrationen in Membrankrümmungen lokalisiert ist (Huang et al., 2006), wie auch die ATP-Synthase-Dimere. Ohne Cardiolipin ist die Rekonstitution nicht geglückt beziehungsweise sind die ATP-Synthase-Dimere weniger gut zueinander angeordnet. Das deutet auf die Wichtigkeit von Cardiolipin in der Stabilisierung der Reihen von ATP-Synthase-Dimeren hin. Weitere Experimente mit verschiedenen ATP-Synthase-Dimeren in verschiedenen Lipiden sind nötig um dies zu untermauern.
Ein weiteres Ziel dieser Arbeit war es ein klonierbares Label zu etablieren, um ein bestimmtes Protein in Kryo-Elektronentomogramme zu identifizieren. Das Label sollte klein sein, um das zu identifizierbare Protein nicht zu beeinflussen und groß genug um in Kryo-Elektronentomogramme identifizierbar zu sein. In Einzelbildern wurde das 6 kDa große Metallothionein gebunden mit Gold identifiziert, wenn zwei Metallothioneine an dem gewünschten Protein kloniert wurden. Metallothionein besteht zu 33 % aus Cysteinen, welche Schwermetalle binden.
In meinen Studien habe ich bewiesen, dass drei Metallothioneine, gebunden mit Gold, in Kryo-Elektronentomogramme detektiert werden können. Jedoch tritt bei der Verwendung von Metallothionein durch die hohe Anzahl an Cysteinen vermehrt Aggregation auf. Bei meinen Untersuchungen fand ich heraus, dass auch das Maltose-Binde-Protein (MBP) ein Signal gleicher Intensität erzeugt. Durch Verwendung von MBP tritt aber keine Aggregation auf und man kann MBP auch zum Reinigen des Proteins verwenden.
Ein früherer Vorschlag zum Reaktionsmechanismus der DFPase, der eine Wasseraktivierung durch den Rest H287 postulierte, mußte nach neuen experimentellen Daten verworfen werden. Daher lag zu Beginn der hier vorliegenden Arbeit kein Vorschlag für den Mechanismus der DFPase vor, der die experimentellen Daten erklären konnte. Für das längerfristige Ziel, die katalytischen Eigenschaften der DFPase gezielt verändern zu können, war es daher notwendig, zunächst den Mechanismus der Hydrolyse von Verbindungen wie DFP durch die DFPase näher zu untersuchen. Durch computergestützte Modellierung im aktiven Zentrum der DFPase (Docking) wurde die Bindung von DFP und anderen Substraten der DFPase genauer untersucht und mit vorhandenen experimentellen Daten verglichen. Diese Arbeiten führten auch zur theoretischen Untersuchung des Bindungsverhaltens von O,O-Dialkylphosphoroamidaten als potentiellen Inhibitoren der DFPase. Diese den Dialkylfluorophosphaten strukturell sehr ähnlichen Substanzen werden durch die DFPase nicht umgesetzt. Die Ergebnisse der Dockinguntersuchung führten schließlich zu der Synthese von drei Phosphoroamidaten unter der Untersuchung ihres Verhaltens als Inhibitoren der DFPase. DIe Charakterisierung erfolgte dabei über enzymkinetische Methoden sowie NMR-Experimente. Mit dem stärksten Inhibitor O,O-Dicyclopentylphosphoroamidat gelang die Kristallisation eines Komlexes mit der DFPase, der mit der Methode der Röntgenbeugung strukturell bestimmt werden konnte. Dieser Komplex belegt die Bindung der Phosphorylgruppe von Substraten an das katalytisch wirksame Calciumion im aktiven Zentrum der DFPase und zeigt die vorherrschenden Bindungskräfte zwischen Ligand und Protein auf. Eine Reihe von Mutanten der DFPase, bei denen gezielt die koordinierenden Aminosäuren des katalytischen Calciumions verändert wurden, ergänzte die bereits in früheren Arbeiten erzeugten Mutanten. Die katalyischen Eigenschaften dieser Mutanten und ihre Fähigkeit zur Metallbindung gestatten es, diese Metallbindungsstelle genauer zu beschreiben und lassen zusammen mit den Dockingexperimenten und der Struktur des Inhibitor-Enzymkomplexes vermuten, dass der calciumkoordinierende Aminosäurerest D229 als aktives Nukleophil im Reaktionsmechanismus der DFPase fungiert. Diese Vermutung ließ sich mit experimentellen Daten untermauern. Durch 18O-Isotopenmarkierung konnte gezeigt werden, dass ein Sauerstoffatom von D229 auf das entstehende Produkt übertragen wird. Hierdurch konnte die Existenz eines Phosphoenzymintermediats nachgewiesen werden. Des weiteren gelang es, Kristalle der DFPase zu züchten, die für Neutronenbeugungsexperimente geeignet waren. Im Rahmen dieser Experimente gelang die Aufnahme eines vollständigen Datensatzes und die Lösung der Neutronenbeugungsstruktur der DFPase in einer Auflösung von 2,2 Å. Diese Neutronenstruktur, in der Wasserstoffatome im Unterschied zu Röntgenstrukturen gut sichtbar sind, zeigt eindeutig, dass der Rest D229 wie erforderlich deprotoniert und dass das in der Bindungstasche an das Calciumion koordinierende Wassermolekül nicht als Hydroxid vorliegt. Damit ließ sich die direkte Aktivierung von Wasser durch das Metallion ausschließen. Neben wichtigen Informationen über die Bindungstasche der DFPase lieferte die Neutronenstruktur auch detaillierte Einblicke in das Wasserstoffbrückennetzwerk im zentralen, wassergefüllten Tunnel des Proteins. Über die Bestimmung des Wasserstoff/Deuteriumaustausches von Proteinrückgradamiden konnten weitere Aussagen über die Solvenszugänglichkeit von verschiedenen Proteinbereichen gemacht werden. Für die DFPase konnten strukturell und funktionell ähnliche Proteine identifiziert werden. Neben der Paraoxonase 1 waren dies das Calciumbindeprotein Regucalcin aus Agrobacterium thumefaciens sowie das Drug Resistance Protein 35 aus Staphylococcus aureus. Es konnte gezeigt werden, dass diese Proteine eine der katalytischen Calciumbindestelle der DFPase vergleichbare Metallbindestelle aufweisen und verschiedene Enzymaktivitäten dieser Proteine durch einen Mechanismus mit einem zu D229 analogen Nukleophil erklärt werden können. Ein direkter Vergleich zwischen der DFPase und der humanen Paraoxonase gelang durch Untersuchungen mit fluorogenen Organophosphaten als Substrate. Hierbei konnte zum ersten Mal gezeigt werden, dass die DFPase in der Lage ist, Substrate mit einer P-O Bindung hydrolytisch zu spalten. Die unterschiedlich gute Umsetzung der verschiedenen Substrate durch die beiden Enzyme konnte auf der Grundlage der Proteinstrukturen erklärt werden, wobei für die Paraoxonase postuliert wurde, dass der calciumbindende Aminosäurerest D269 als zu D229 analoges Nukleophil wirkt. Abschließend gelang es, zusätzlich eine Zusammensetzung für eine Enzympräparation zu finden, die eine Gefriertrocknung der DFPase mit nur geringem Verlust an enzymatischer Aktivität erlaubt. Ein solches lagerstabiles Enzympulver ist ein notwendiger Schritt hin zu einer praktischen Anwendung der DFPase für die technische Dekontamination von hochtoxischen Nervenkampfstoffen.
The translation eukaryotic elongation factor 1alpha (eEF1A) is a monomeric GTPase involved in protein synthesis. In addition, this protein is thought to participate in other cellular functions such as actin bundling, cell cycle regulation, and apoptosis. Here we show that eEF1A is associated with the alpha2 subunit of the inhibitory glycine receptor in pulldown experiments with rat brain extracts. Moreover, additional proteins involved in translation like ribosomal S6 protein and p70 ribosomal S6 protein kinase as well as ERK1/2 and calcineurin were identified in the same pulldown approaches. Glycine receptor activation in spinal cord neurons cultured for 1 week resulted in an increased phosphorylation of ribosomal S6 protein. Immunocytochemistry showed that eEF1A and ribosomal S6 protein are localized in the soma, dendrites, and at synapses of cultured hippocampal and spinal cord neurons. Consistent with our biochemical data, immunoreactivities of both proteins were partially overlapping with glycine receptor immunoreactivity in cultured spinal cord and hippocampal neurons. After 5 weeks in culture, eEF1A immunoreactivity was redistributed to the cytoskeleton in about 45% of neurons. Interestingly, the degree of redistribution could be increased at earlier stages of in vitro differentiation by inhibition of either the ERK1/2 pathway or glycine receptors and simultaneous N-methyl-D-aspartate receptor activation. Our findings suggest a functional coupling of eEF1A with both inhibitory and excitatory receptors, possibly involving the ERK-signaling pathway.
The theoretical IR-frequencies of sulphurdifluoride are computed from force constants, which are evaluated by means of molecules with S-F-bonds: ν1 = 795 ± 10cm-1, ν2 = 430± 5cm-1, ν3= 830 ± 10cm-1.
Der Einsatz von Mikrowellen zur Synthese von Organosilicioumverbindungen ist bislang nicht beschrieben und wird mit der vorliegenden Arbeit eingeführt. Dazu wurde eine Haushaltsmikrowelle durch Öffnen des Sicherheitskäfigs und entsprechender Abschirmung so modifiziert, dass mit den üblichen Labormaterialien gearbeitet werden konnte. Exemplarische Reaktionen in flüssiger Phase, wie die Synthese von Silatranen und silatrananalogen Verbindungen, können gegenüber der klassischen Reaktionsführung bis zum Faktor 60 bei vergleichbaren Ausbeuten und Reinheiten beschleunigt werden. Auch die direkte Konversion von SiO2 in reaktive Verbindungen gelingt unter Mikrowellenbedingungen deutlich beschleunigt. Weitere Modifikationen der Apparatur ermöglichen die Durchführung von Festphasen-Gas Reaktionen unter Verwendung von Silicium und verschiedenen Reaktionsgasen. Verwendet wurden dazu Cl2, HCl, CH3Cl sowie Gemishce dieser Gase. Auch wurden die Reaktionen unter verschiedenen Stufen der Argonverdünnung durchgeführt. Erstaunlicherweise glüht dabei das Silicium innerhalb von Sekunden mit einer Temperatur von über 1000°C. Die Untersuchungen zeigen eine hohe Tendenz zur Bildung von monomeren Siliciumverbindungen, sobald Methylchlorid beteiligt ist, wird bevorzugt das technisch bedeutsame Me2SiCl2 beobachtet. Alle Ergebnisse gehen konform mit der Annahme, dass durche Mikrowelle aktiviertes Silicium bereitgestellt wird. Dadurch kann es zur Bildung und Stabilisierung von intermediären Silylenen kommen, die in verschiedene Bindungen der Reaktionspartner insertieren. Bemerkenswert ist auch die geringe elektrische Leistung von 200 Watt, die in allen diskutierten Umsetzungen ausreicht, um die gewünschten Reaktionen durchzuführen. Darüber hinaus wurde auch der Unterschied zwischen Multimode- und Singlemodegeräten untersucht. Nur bei Verwendung von Multimodegeräten könenn die beschriebenen Ergebnisse erzielt werden. Beim Einsatz von Singlemodegeräten entsprechen die Resultate denen, die bei klassischer thermischer Reaktionsführung erzielt werden.
Loss of the tumor suppressor Pdcd4 was reported for various tumor entities and proposed as a prognostic marker in tumorigenesis. We previously characterized decreased Pdcd4 protein stability in response to mitogenic stimuli, which resulted from p70S6K1-dependent protein phosphorylation, β-TrCP1-mediated ubiquitination, and proteasomal destruction. Following high-throughput screening of natural product extract libraries using a luciferase-based reporter assay to monitor phosphorylation-dependent proteasomal degradation of the tumor suppressor Pdcd4, we succeeded in showing that a crude extract from Eriophyllum lanatum stabilized Pdcd4 from TPA-induced degradation. Erioflorin was identified as the active component and inhibited not only degradation of the Pdcd4-luciferase-based reporter but also of endogenous Pdcd4 at low micromolar concentrations. Mechanistically, erioflorin interfered with the interaction between the E3-ubiquitin ligase β-TrCP1 and Pdcd4 in cell culture and in in vitro binding assays, consequently decreasing ubiquitination and degradation of Pdcd4. Interestingly, while erioflorin stabilized additional β-TrCP-targets (such as IκBα and β-catenin), it did not prevent the degradation of targets of other E3-ubiquitin ligases such as p21 (a Skp2-target) and HIF-1α (a pVHL-target), implying selectivity for β-TrCP. Moreover, erioflorin inhibited the tumor-associated activity of known Pdcd4- and IκBα-regulated αtranscription factors, that is, AP-1 and NF-κB, altered cell cycle progression and suppressed proliferation of various cancer cell lines. Our studies succeeded in identifying erioflorin as a novel Pdcd4 stabilizer that inhibits the interaction of Pdcd4 with the E3-ubiquitin ligase β-TrCP1. Inhibition of E3-ligase/target-protein interactions may offer the possibility to target degradation of specific proteins only as compared to general proteasome inhibition.
The tumor suppressor programmed cell death 4 (Pdcd4) exerts its function by inhibiting protein translation initiation. Specifically, it displaces the scaffold protein eukaryotic initiation factor 4G (eIF4G) from its binding to the eukaryotic initiation factor 4A (eIF4A). Thereby, Pdcd4 inhibits the helicase activity of eIF4A, which is necessary for the unwinding of highly structured 5’ untranslated regions (UTRs) of messenger RNAs (mRNAs) often found in oncogenes like c-myc to make them accessible for the translation machinery and subsequent protein production. Overexpression of Pdcd4 inhibits tumorigenesis in vitro and in vivo and inversely, Pdcd4 knockout mice show enhanced tumor formation. In line, Pdcd4 is lost in various tumor types and proposed as prognostic factor in colon carcinomas. Unlike most other tumor suppressors that are rendered nonfunctional by mutations (e.g., p53), Pdcd4 loss is not attributable to mutational inactivation. It is regulated via translational repression by microRNAs and increased degradation of the protein under tumor promoting, inflammatory conditions and mitogens. Specifically, proteasomal degradation of Pdcd4 is controlled by p70 S6 Kinase (p70S6K)-mediated phosphorylation in its degron sequence (serines 67, 71 and 76). Stimulation of the PI3K-AKT-mTOR pathway by growth factors, hormones and cytokines initiates p70S6K activity. Phosphorylated Pdcd4 is subsequently recognized by the E3 ubiquitin ligase beta-transducin repeats-containing protein (β-TrCP) and marked with a polyubiquitin tail to be detected by the 26S proteasome for degradation. β-TrCP represents the substrate specific recognition subunit of the ubiquitin ligase complex responsible for protein-protein interaction with Pdcd4 as substrate for ubiquitin transfer and subsequent proteasomal disassembly.
The first part of the present work aimed at identifying novel stabilizers of the tumor suppressor Pdcd4 in a high throughput screen (HTS). As assay design, a fragment of Pdcd4 from amino acid 39 to 91, containing the phosphorylation sensitive degron sequence, was fused to a luciferase reporter gene construct. Stable expression of this Pdcd4(39-91)luciferase (Pdcd4(39-91)luc) fusion protein in HEK 293 cells served as read-out for the Pdcd4 protein amount to be detected in a high throughput compatible cell-based assay. Loss of Pdcd4(39-91)luc was induced by treatment with 12-O-
tetradecanoylphorbol-13-acetate (TPA), a phorbolester, which activates the PI3K signaling cascade leading to degradation of Pdcd4. The cut-off for hit definition was set at >50% activity in rescuing the Pdcd4(39-91)luc signal from TPA-induced degradation. Activity was calculated relative to the difference of DMSO- and TPA-treated cells (ΔDMSO-TPA = RLUDMSO-RLUTPA). Initial screening of a protein kinase inhibitor library (PKI) revealed hit substances expected to show Pdcd4 stabilizing activity by inhibition of kinases involved in Pdcd4 downregulation, e.g., the mTOR inhibitor rapamycin, the PI3K inhibitors wortmannin and LY294002 and the PKC inhibitors GF 109203X and Ro 31-8220.
The Molecular Targets Laboratory (MTL) of the National Cancer Institute (NCI) in Frederick, USA, hosts one of the largest collections of crude natural product extracts as well as a big substance libraries from pure synthetic sources. Screening of over 15 000 pure compounds and over 135 000 natural product extracts identified 46 pure and 42 extract hits as Pdcd4 stabilizers. For nine synthetic and six natural product derived compounds (after bioassay-guided fractionation), dose-dependent activities for recovering the TPA-induced Pdcd4(39-91)luc loss defined IC50s in the low micromolar range. Most importantly, these compounds were confirmed to stabilize endogenous Pdcd4 protein levels from forced degradation as well. This result proved the assay design to be highly representative for endogenous cellular mechanisms regulating Pdcd4 protein stability. The next step was to stratify the hit substances according to their likely mechanism of action to be located either up- or downstream of the p70S6K-mediated phosphorylation of Pdcd4. Therefore, phosphorylation of S6, as proto-typical p70S6K target, was analyzed and uncovered two natural derived compounds to influence p70S6K activity. Four substances did not affect p70S6K phosphorylation activity and were therefore considered to stabilize Pdcd4 by acting downstream, i.e. on the β-TrCP-mediated proteasomal degradation.
In the second part of this work, one of these compounds, namely the sesquiterpene lactone erioflorin, isolated by bioassay-guided fraction from the active extract of Eriophyllum lanatum, Asteraceae, was further characterized in detail with respect to its molecular mechanism of action. Erioflorin dose-dependently protected both Pdcd4(39-91)luc and endogenous Pdcd4 protein from TPA-induced degradation with IC50s of 1.28 and 2.64 μM, respectively. Pdcd4 stabilizing activity was maximal at 5 μM erioflorin. Up to this concentration, erioflorin was verified not to inhibit p70S6K activity. In addition, it was observed that erioflorin rescued Pdcd4(39-91)luc from both, wild type and constitutively active p70S6K-mediated downregulation. Only wild type p70S6K was inhibitable by the mTOR inhibitor rapamycin which served as an upstream acting control. To study the next section of Pdcd4 regulation, i.e. recognition by the E3 ubiquitin ligase β-TrCP, Pdcd4(39-91)luc and endogenous Pdcd4 were immunoprecipitated from whole cell extracts with the corresponding antibodies. In this key experiment, treatment with TPA increased overexpressed β-TrCP binding to both and this coimmunoprecipitation could be strongly reduced by erioflorin treatment. This result strongly pointed to an inhibitory mechanism of the β-TrCP specific binding to Pdcd4 by erioflorin. In addition, erioflorin disrupted the binding of in vitro transcribed/translated β-TrCP to Pdcd4 in an in vitro interaction assay to exclude nonspecific intracellular signals. Furthermore, polyubiquitination of Pdcd4 was decreased by erioflorin treatment as well. To clarify questions regarding specificity of erioflorin for the E3 ubiquitin ligase β-TrCP, stability of another important β-TrCP target was explored, i.e. the tumor suppressor inhibitor of kappa B alpha (IκBα). Indeed, the tumor necrosis factor alpha (TNFα)-mediated loss of IκBα could be prevented by erioflorin cotreatment. On the other hand, the E3 ubiquitin ligase von Hippel Lindau protein (pVHL) was left unaffected as its target hypoxia inducible factor 1 alpha (HIF-1α) could not be stabilized from oxygen-dependent degradation by erioflorin treatment. These results argued strongly for erioflorin being a specific inhibitor of β-TrCP-mediated protein degradation. Functional consequences of erioflorin treatment were investigated by observing its influence on the transcriptional activities of the transformation marker activator protein 1 (AP-1, an indirect downstream target of Pdcd4) and nuclear factor κB (NF-κB which is directly inhibited by IκBα). Indeed, erioflorin showed significant inhibition of AP-1 and NF-κB reporter constructs at 5 μM, a concentration for which an impact on cell viability was excluded. Finally to characterize the significance of erioflorin in a cell-based tumorigenesis assay, the highly invasive colon carcinoma cell line RKO was tested in a two dimensional migration assay. Erioflorin was discovered to significantly lower cell migration in a wound closure assay.
In conclusion, development of a high throughput compatible cell-based reporter assay successfully identified novel substances from pure synthetic and natural product derived background as potent stabilizers of the tumor suppressor Pdcd4. In addition, this work aimed at elucidating the detailed mechanism of action of the sesquiterpene lactone erioflorin from Eriophyllum lanatum, Asteraceae. Erioflorin was discovered to inhibit the E3 ubiquitin ligase β-TrCP, thereby preventing protein degradation of tumor suppressors like Pdcd4 and IκBα. This may offer the possibility to more specifically target protein degradation and generate less adverse side effects by blocking a particular E3 ubiquitin ligase compared to general proteasome inhibition.
The title compound, [Tl4(C4H9O)4], featuring a (Tl—O)4 cube, crystallizes with a quarter-molecule (located on a special position of site symmetry An external file that holds a picture, illustration, etc. Object name is e-66-m1621-efi1.jpg..) and a half-molecule (located on a special position of site symmetry 23.) in the asymmetric unit. The Tl—O bond distances range from 2.463 (12) to 2.506 (12) Å. All O—Tl—O bond angles are smaller than 90° whereas the Tl—O—Tl angles are wider than a rectangular angle.
The title compound, C25H20N4O2, is a ditopic ortho-hydroquinone-based bis(pyrazol-1-yl)methane ligand. The dihedral angles between the planes of the pyrazole rings and their attached phenyl rings are 17.4 (3) and 5.9 (4)°. The pyrazole rings make a dihedral angle of 87.84 (16)°. One of the two hydroxy groups forms an intramolecular hydrogen bond to the other hydroxy group, whereas the second is involved in an intermolecular O—H[cdots, three dots, centered]N hydrogen bond. As a result of these intermolecular hydrogen bonds, helical chains running along the b axis are formed.
The IrIII atom of the title compound, [Ir(C11H8N)2Cl(CH3CN)], displays a distorted octahedral coordination. The pyridyl groups are in trans positions [N—Ir—N = 173.07 (10)°], while the phenyl groups are trans with respect to the acetonitrile and chloride groups [C—Ir—N = 178.13 (11) and C—Ir—Cl = 176.22 (9)°]. The pyridylphenyl groups only show a small deviation from planarity, with the dihedral angle between the planes of the two six-membered rings in each pyridylphenyl group being 5.6 (2) and 5.8 (1)°. The crystal packing shows intermolecular C—H[cdots, three dots, centered]Cl, C—H[cdots, three dots, centered]π(acetonitrile) and C—H[cdots, three dots, centered]π(pyridylphenyl) contacts.
Die vorliegende Arbeit beschäftigte sich mit der Entwicklung, Synthese und
Charakterisierung neuartiger redoxaktiver Liganden und deren Metallkomplexen. Basierend
auf dem para- und ortho-Hydrochinon / Benzochinon-Redoxsystem wurden 13 neue
Bis(pyrazol-1-yl)methan-Liganden (28 – 36 und 67 – 70; Schema 47) synthetisiert und
vollständig charakterisiert. Ein Schwerpunkt lag auf der Einführung von Substituenten am
Bis(pyrazol-1-yl)methan-Donor, um deren Einfluss auf das N,N′-Koordinationsverhalten
gegenüber Metallionen zu untersuchen. In Analogie zu den klassichen Skorpionaten sind
Substituenten in Position 3 der Pyrazolringe in der Lage, koordinativ ungesättigte
Metallzentren kinetisch zu stabilisieren, was für potentielle Anwendungen in der Katalyse
essentiell ist. Bei den ortho-chinoiden Liganden (67 – 70; Schema 47) erfüllt die redoxaktive
Gruppe eine zweite Funktion, nämlich als Chelatdonor gegenüber Metallzentren, was die
Synthese und Untersuchung (hetero-)dinuklearer Komplexe erlaubt.

Die kristallographische Charakterisierung von 10 dieser Liganden (28 – 33, 67 – 70) zeigte
größtenteils sehr ähnliche strukturelle Parameter. Ein steigender sterischer Anspruch der
Substituenten am Bis(pyrazol-1-yl)methan führte zu einer leichten Streckung der
Chinon–Bis(pyrazol-1-yl)methan-Bindung und zu kurzen Kontakten zwischen Substituenten
am zentralen Methin-Kohlenstoffatom und den ipso- (HQ-C1) bzw. ortho-Kohlenstoffatomen
(HQ-C2) am sechsgliedrigen Ring. Diese kurzen Kontakte spielten in der oxidativen
Demethylierung von 32 eine Rolle. Während alle anderen para-chinoiden Liganden mit
Cerammoniumnitrat (CAN) zu den erwarteten para-Benzochinon-Derivaten reagierten
(Schema 48), wurde im Zuge der Oxidation von 32 ein zusätzliches Sauerstoffatom am
sechsgliedrigen Ring eingeführt (47; Schema 48). Im Gegenzug wurde die sterisch am
stärksten abgeschirmte Methoxygruppe nicht oxidativ demethyliert. Letztendlich konnte
gezeigt werden, dass (i) das neu eingeführte Sauerstoffatom von atmosphärischem Sauerstoff
stammt und (ii) alle fünf Methylgruppen und beide Methoxygruppen in 32 für die Oxidation
essentiell sind.

Die Cyclovoltammogramme der ortho-chinoiden Bis(pyrazol-1-yl)methan-Liganden
(untersucht am Beispiel von 67, 68 und 70) zeigten irreversible Redoxwellen, da im Zuge der
Oxidation OH-Protonen abgespalten wurden; die Redoxpotentiale liegen in einem mit
chemischen Oxidationsmitteln gut zugänglichen Bereich. 70 wurde von CAN erfolgreich
oxidiert, das Produkt 71 zersetzte sich unter den Reaktionsbedingungen allerdings sehr
schnell und konnte nicht isoliert, sondern lediglich als Additionsprodukt von 4-tert-
Butylpyridin abgefangen werden. Unter optimierten Reaktionsbedingungen und mit DDQ als
Oxidationsmittel ließen sich 70 und 68 in ihre oxidierte Form überführen und in Reinform
gewinnen. Die für ortho-Benzochinone typische Neigung zur Zersetzung wurde auch bei 71
und 73 beobachtet, wobei letzteres sich wesentlich schneller zersetzte (innerhalb von
Stunden) als 71 (innerhalb eines Tages).

Die Koordinationschemie repräsentativer Vertreter der 13 redoxaktiven Bis(pyrazol-1-
yl)methan-Liganden wurde untersucht. Bereits der sterisch nur mäßig anspruchsvolle parachinoide
Ligand 29 ist in der Lage, koordinativ ungesättigte CoII-Ionen kinetisch gegenüber
der Bildung von 1:2 Komplexen zu stabilisieren. Im Festkörper liegen ausschließlich
Verbindungen mit einer 1:1 Zusammensetzung von Ligand zu CoII vor (59 und 60; Abb. 36).
In Lösung scheinen hingegen Gleichgewichte zu existieren, in denen auch die koordinativ
abgesättigten oktaedrischen 2:1 Komplexe auftreten. Die ortho-chinoiden Liganden 67 und 68
bildeten selektiv entsprechende N,N′-koordinierte PdCl2-Komplexe, ohne dass das ortho-
Hydrochinonat (Catecholat) als konkurrierender O,O′-Donor wirkte (74 und 75).

Es zeigte sich jedoch auch, dass sterisch sehr anspruchsvolle Substituenten am
Bis(pyrazol-1-yl)methan-Fragment in Reaktionen mit Übergangsmetallen zu einer Zersetzung
des Ligandengerüsts führen können. So reagierte der para-chinoide tert-Butyl-substituierte
Ligand 31 mit [Co(NO3)2] zu [(HpztBu,H)2Co(NO3)2] (63). Eine analoge Zersetzung zu trans-
[(HpzR,H)2PdCl2] (76: R = Ph und 77: R = tBu) wurde nach der Reaktion der ortho-chinoiden
Liganden 69 bzw. 70 mit [PdCl2]-Quellen beobachtet.

Die ortho-chinoiden Bis(pyrazol-1-yl)methan-Liganden (67 – 70) besitzen mit ihrem
Catecholat-O,O′-Donor eine zweite Koordinationsstelle, was diese Liganden für die Synthese
von dinuklearen Komplexen interessant macht. Da gezeigt werden konnte, dass [PdCl2]
selektiv an den Bis(pyrazol-1-yl)methan-Donor koordiniert (vgl. 59, 60, 74 und 75; Abb. 36),
galt es als nächstes zu evaluieren, ob eine ähnlich selektive Bindung anderer Metallionen an
den O,O′-Donor möglich ist.

In der Tat konnten in sehr guten Ausbeuten O,O′-gebundene [(p-cym)Ru]-, [(Phpy)2Ir]-
und [(Cp*)Ir]-Komplexe ausgewählter redoxaktiver ortho-chinoider Liganden dargestellt
werden. Vorteilhaft war die Verwendung der kristallinen, nicht-flüchtigen Base TlOtBu zum
Abfangen der im Zuge der Komplexierung freiwerdenden Protonen (Schema 49, Abb. 37).
Die Eliminierung von TlCl sorgt für eine irreversible Reaktion zu den entsprechenden

Komplexen. Besonders interessant ist die Koordinationschemie des Liganden 68 im chiralen,
anionischen IrIII-Komplex 83 (Abb. 37 Mitte), der in der Synthese als Thallium-Salz anfiel
und im Festkörper TlI-verbrückte Dimere bildete.
Eine elektrochemische Charakterisierung wurde mit 85 durchgeführt. Wie erwartet, zeigte
der komplexierte Bis(dimethylpyrazol-1-yl)methan-Ligand im Gegensatz zu freiem 68 eine
reversible Oxidationswelle. Die Potentialdifferenz zwischen Liganden-Oxidation und Iridium-
Reduktion beträgt fast 2 V, was in diesem Zusammenhang bedeutet, dass sich 68 als
unschuldiger Ligand verhält und man Iridium zweifelsfrei die Oxidationsstufe +III zuweisen
kann. Mit Komplexen von 68 und leichter reduzierbaren Übergangsmetallen sollte es
hingegen möglich sein, in den Bereich des nicht-unschuldigen Verhaltens vorzudringen und
z.B. Valenz-Tautomerie zu beobachten.
Mit effizienten Synthesewegen zu N,N′-Komplexen einerseits (74 und 75; Abb. 36) und
O,O′-Komplexen andererseits (u.a. 83 und 85; Abb. 37) wurde als nächstes ein heterodinuklearer
Komplex synthetisiert (87; Schema 50). 68 erwies sich als am besten geeigneter
Ligand, da er, wie bereits erwähnt, eine gute Löslichkeit bei moderatem sterischen Anspruch
besitzt und der N,N′-Donor auch in Gegenwart von Lewis-sauren Metallionen beständig ist.
Die Wannenform des Bis(pyrazol-1-yl)methan-PdII-Chelatrings in 87 bringt das Palladium-
Ion in räumliche Nähe zum ortho-Hydrochinonat π-System. In früheren Studien dieser
Arbeitsgruppe konnte gezeigt werden, dass ein solches Arrangement Elektronenübertragungen
zwischen dem Chinon und dem koordinierten Palladium-Zentrum erlaubt.
Durch die direkte konjugative Wechselwirkung des zweiten Metallzentrums (IrIII) mit der
redoxaktiven Gruppe über die Sauerstoffatome in 87 sollte eine effektive elektronische
Ligand ↔ Metall- und Metall ↔ Metall-Kommunikation möglich sein. Die elektrochemische
Charakterisierung zeigte allerdings, dass im vorliegenden Fall die Potentiale der drei
Komponenten Chinon / PdII / IrIII mit jeweils ca. einem Volt zu weit auseinander liegen.

und 70 für den Aufbau höhermolekularer Koordinationsverbindungen und oligonuklearer

Dafür wurden Komplexliganden synthetisiert, die aus einem Zentralmetall bestehen, das
oktaedrisch von je drei O,O′-koordinierenden Liganden 67 bzw. 70 umgeben ist (Schema 51).
Mit den freien Bis(pyrazol-1-yl)methan-Donorgruppen vermag jeder dieser Komplexliganden
drei weitere Metallzentren zu koordinieren. Derartige Verbindungen könnten aufgrund ihrer
dreidimensionalen Struktur z.B. Anwendung im Aufbau von redoxaktiven metallorganischen
Netzwerken oder elektrisch leitfähigen Koordinationspolymeren finden. Als Zentralmetalle
dienten FeIII- und AlIII-Ionen; erstere, weil sie selbst redoxaktiv sind, und letztere, weil sie
eine im Vergleich zu FeIII ähnliche Koordinationschemie besitzen, aber wegen ihres
Diamagnetismus‘ eine NMR-spektroskopische Charakterisierung ermöglichen. Je nach
verwendeter Base wurden die dreifach anionischen Komplexliganden als Lithium- bzw.
Thallium-Salze isoliert. Die NMR-Spektren von 89, 90, 93 und 94 zeigten jeweils nur einen
Signalsatz, obwohl sich, bedingt durch die Asymmetrie des Liganden und die Chiralität des
oktaedrischen Metallzentrums, fac- und mer-Isomere bilden können. Es konnte nicht
abschließend geklärt werden, ob sich exklusiv das höhersymmetrische fac-Isomer bildet, oder
ob ein Mechanismus aktiv ist, der alle Isomere auf der NMR-Zeitskala schnell ineinander
überführt, sodass die Gegenwart lediglich einer Spezies vorgetäuscht wird. In
elektrochemischen Untersuchungen zeigten die Komplexliganden mehrere irreversible
Oxidationswellen. Ob dieses Verhalten auf eine intramolekulare Kommunikation der
einzelnen redoxaktiven Gruppen zurückzuführen ist, müssen weitergehende Studien zeigen.
Als prinzipieller Beleg für die präparative Anwendbarkeit der Komplexliganden und
Grundlage für die Untersuchung der Koordinationschemie der oktaedrischen
Komplexliganden, wurden PdII-Komplexe von 89 und 93 synthetisiert.

Although the constituents of the SD are largely known, its molecular organization remains elusive. Here, we use super-resolution correlative light and electron microscopy to quantify a linear rate of reduction in albumin concentration across the filtration barrier. Next, we use cryo-electron tomography of vitreous lamellae from high-pressure frozen native glomeruli to analyze the molecular architecture of the SD. The resulting densities resemble a fishnet pattern. Fitting of Nephrin and Neph1, the main constituents of the SD, results in a complex interaction pattern with multiple contact sites between the molecules. Using molecular dynamics flexible fitting, we construct a blueprint of the SD, where we describe all interactions. Our architectural understanding of the SD reconciles previous findings and provides a mechanistic framework for the development of novel therapies to treat kidney dysfunction.

We present the rapid biophysical characterization of six previously reported putative G‐quadruplex‐forming RNAs from the 5′‐untranslated region (5′‐UTR) of silvestrol‐sensitive transcripts for investigation of their secondary structures. By NMR and CD spectroscopic analysis, we found that only a single sequence—[AGG]2[CGG]2C—folds into a single well‐defined G‐quadruplex structure. Sequences with longer poly‐G strands form unspecific aggregates, whereas CGG‐repeat‐containing sequences exhibit a temperature‐dependent equilibrium between a hairpin and a G‐quadruplex structure. The applied experimental strategy is fast and provides robust readout for G‐quadruplex‐forming capacities of RNA oligomers.
Natürliche Enantioselektivität und Isotopendiskriminierung - Schlüssel zur Echtheit ätherischer Öle

Als Grundlage für die Beurteilung der Echtheit ätherischer Öle können zwei biochemische Prinzipien Enantioselektivität und Isotopendiskriminierung während der Biosynthese herangezogen werden. In der vorliegenden Arbeit wurde die enantioselektive Kapillargaschromatographie sowie die online-Kopplung der Gaschromatographie mit der Isotopenmas- senspektrometrie zur Authentizitätsbewertung verschiedener ätherischer Öle eingesetzt. Die Bestimmung von Enantiomerenverhältnissen mittels Multidimensionaler Gaschromatographie-Massenspektrometrie (MDGC-MS) sowie von 13C/12C-Isotopenverhältnissen mittels GC-C-IRMS (Gaschromatographie-Combustion- Isotopenmassenspektrometrie) sind etablierte Methoden, die in der Authentizitätsbewertung von Aroma- und Duftstoffen eingesetzt werden. Dagegen ist die Bestimmung von 2H/1H-Isotopenverhältnissen mittels GC-P-IRMS (Gaschromatographie-Pyrolyse-Isotopenmassenspektrometrie) eine relativ neue Methode. In der vorliegenden Arbeit wurden Strategien zur Bestimmung von zuverlässigen 2H/1H-Isotopenverhältnissen mittels GC-P-IRMS entwickelt. Die Kalibrierung des Referenzgases mit Hilfe von internationalen Standards kann nur mittels eines Elemental Analyzers (EA-IRMS) erfolgen, da für die Gaschromatographie geeignete Standards nicht zur Verfügung stehen. Daher ist insbesondere der Vergleich von Isotopenverhältnissen von Standardsubstanzen, die mittels TC/EA-IRMS und GC- P-IRMS bestimmt wurden, von Bedeutung. Es konnte gezeigt werden, dass eine Konditionierung (Einbringen einer Kohlenstoffschicht) des Pyrolysereaktors im GC- P-IRMS-System notwendig ist, um für die untersuchten Aromastoffe mittels GC-P- IRMS zum Elemental Analyzer vergleichbare Ergebnisse zu erzielen. Von zwei verschiedenen getesteten Methoden zur Konditionierung des Pyrolysereaktors war die Konditionierung durch Einleiten von Methan in den Pyrolysereaktor die geeignetere und effektivere Methode. Weiterhin wurden der Einfluss des Trägergasflusses des Gaschromatographen auf die bestimmten Isotopenwerte sowie der lineare Bereich der Methode untersucht. Es konnte erstmalig gezeigt werden, dass die mittels GC-P-IRMS bestimmten Isotopenverhältnisse von verschiedenen Parametern abhängig sind. Richtige Ergebnisse zu erzielen setzt folgende Konditionen voraus: Konditionierung des Pyrolysereaktors, optimaler Trägergasfluss sowie Mindestmenge des Analyten (> 0,3 µg on column). Die neuen Möglichkeiten, die die online-Bestimmung von 2H/1H-Isotopenverhältnissen in der Authentizitätsbewertung von Lavendelölen, Anis- und Fenchelölen sowie Kümmelölen bietet, wurden erstmalig untersucht. Darüber hinaus wurden die enantioselektive Kapillargaschromatographie und die Bestimmung von 13C/12C-Isotopenverhältnissen mittels GC-C-IRMS eingesetzt. Es konnte gezeigt werden, dass sich die Bereiche der 2HV-SMOW-Werte von Linalool und Linalylacetat aus authentischen Lavendelölen deutlich vom Bereich der Isotopenverhältnisse kommerziell erhältlicher, synthetischer Analoga unterscheiden und somit eine Verfälschung von Lavendelölen mit synthetischem Linalool und/oder Linalylacetat nachweisbar ist. Mit dieser Methode konnten diverse Handelsöle eindeutig als verfälscht beurteilt werden. Über die Bestimmung der Enantiomerenverhältnisse von Linalool und Linalylacetat dieser Öle konnte die Aussage bestätigt werden (hohe Reinheit zugunsten des (R)-Enantiomeren in genuinen Lavendelölen). Anhand der unterschiedlichen 13C/12C-Isotopenverhältnisse ist eine Unterscheidung zwischen synthetischem und Linalylacetat aus Lavendel möglich. Die 13CV-PDB-Werte von synthetischem und natürlichem Linalool aus Lavendelölen liegen im gleichen Bereich und sind somit zur analytischen Differenzierung natürlich/naturidentisch nicht geeignet. Mittels GC-C(P)-IRMS Analyse von trans-Anethol aus Fenchel- und Anisölen wurden die authentischen Bereiche der 13C/12C- und 2H/1H-Isotopenverhältnisse ermittelt. Es konnte gezeigt werden, dass sich einige der synthetischen trans-Anethol- Muster nur aufgrund eines der beiden bestimmten Isotopenverhältnisse von dem authentischen Bereich abgrenzen lassen. Somit konnte gezeigt werden, dass die integrale Betrachtungsweise der 13CV-PDB-Werte und der 2HV-SMOW-Werte biogener Stoffe für die Authentizitätsbewertung von großer Bedeutung ist. Zur Authentizitätsbewertung von Kümmelölen wurden die Enantiomerenverhältnisse, die 13C/12C- und die 2H/1H-Isotopenverhältnisse der Hauptkomponenten Limonen und Carvon bestimmt. Aufgrund der 2HV-SMOW-Werte von Limonen lassen sich keine Unterscheidungen zwischen kommerziell erhältlichen Limonen und Limonen aus Kümmelölen treffen. Dagegen können die 13CV-PDB-Werte von Limonen zur Authentizitätsbewertung von Kümmelölen herangezogen werden. Die dargestellten Ergebnisse zeigen, dass die Bestimmung von 2H/1H- Isotopenverhältnissen mittels GC-P-IRMS neue Möglichkeiten in der Echtheitsbewertung ätherischer Öle bietet. Insbesondere die integrale Betrachtung von 2HV- SMOW- und 13CV-PDB-Werten ( 2H/ 13C-Korrelation) wird eine Authentizitätsbewertung künftig noch wesentlich differenzierter möglich machen. Weitere Perspektiven wird die Bestimmung von 18O/16O-Isotopenverhältnissen bieten. Die Authentizitätsbewertung anhand der Multielement-Analyse mittels GC- IRMS eröffnet gerade für achirale Aromastoffe Perspektiven, stellt aber auch eine Ergänzung zur enantioselektiven Analytik dar.
By adopting a variety of shapes, proteins can perform a wide number of functions in the cell, from being structural elements or enabling communication with the environment to performing complex enzymatic reactions needed to sustain metabolism. The number of proteins in the cell is limited by the number of genes encoding them. However, several mechanisms exist to increase the overall number of protein functions. One of them are post-translational modifications, i.e. covalent attachment of various molecules onto proteins. Ubiquitin was the first protein to be found to modify other proteins, and, faithful to its evocative name, it is involved in nearly all the activities of a cell. Ubiquitylation of proteins was believed for a long time only to be responsible for proteasomal degradation of modified proteins. However, with the discovery of various types of ubiquitylation, such as mono-, multiple- or poly-ubiquitylation, new functions of this post-translational modification emerged. Mono-ubiquitylation has been implicated in endocytosis, chromatin remodelling and DNA repair, while poly-ubiquitylation influences the half-life of proteins or modulates signal transduction pathways. DNA damage repair and tolerance are example of pathways extensively regulated by ubiquitylation. PCNA, a protein involved in nearly all types of DNA transaction, can undergo both mono- and poly-ubiquitylation. These modifications are believed to change the spectrum of proteins that interact with PCNA. Monoubiquitylation of PCNA is induced by stalling of replication forks when replicative polymerases (pols) encounter an obstacle, such as DNA damage or tight DNA-protein complexes. It is believed that monoubiquitylation of PCNA stimulates the exchange between replicative pols to one of polymerases that can synthesize DNA across various lesions, a mechanism of damage tolerance known as translesion synthesis (TLS). Our work has helped to understand why monoubiqutylation of PCNA favours this polymerase switch. We have identified two novel domains with the ability to bind Ub non-covalently. These domains are present in all the members of Y polymerases performing TLS, and were named Ub-binding zinc finger (UBZ) (in polη and polκ) and Ub-binding motif (UBM) (in polι and Rev1). We have shown that these domains enable Y polymerases to preferentially gain access to PCNA upon stalling of replication, when the action of translesion polymerases is required. While the region of direct interaction between Y pols and PCNA had been known (BRCT domain in Rev1 and PIP box motif (PIP) in three others members), we propose that Ub-binding domains (UBDs) in translesion Y pols enhance the PIP- or BRCT-domain-mediated interaction between these polymerases and PCNA by binding to the Ub moiety attached onto PCNA. Following these initial studies, we have also discovered that Y polymerases themselves undergo monoubiquitylation and that their UBDs mediate this modification. This auto-ubiquitylation is believed to lead to an intramolecular interaction between UBD and Ub attached in cis onto the UBD-containing protein. We have mapped monoubiquitylation sites in polη in the C-terminal portion of the protein containing the nuclear localization signal (NLS) and the PIP box. Beside PIP, the NLS motif is also involved in direct interaction of polη with PCNA. Based on these findings, we propose that monoubiquitylation of either NLS or PIP masks them from potential interaction with PCNA. Lastly, using several functional assays, we have demonstrated the importance of all these three motifs in the C-terminus of polη (UBZ, NLS and PIP) for efficient TLS. We have also constructed a mimic of monoubiquitylated polη by genetically fusing polη with Ub. Interestingly, this chimera is deficient in TLS as compared to the wild-type protein. Altogether, these studies demonstrate that the C-terminus of polη constitutes a regulatory module involved in multiple-site interaction with monoubiquitylated PCNA, and that monoubiquitylation of this region inhibits the interaction between polη and PCNA. Our work has also revealed that the UBDs of Y pols as well as of other proteins implicated in DNA damage repair and tolerance, such as the Werner helicase-interacting protein 1 (Wrnip1), are required for their proper sub-nuclear localization. All these proteins localize to discrete focal structures inside the nucleus and mutation of their UBDs results in inability to accumulate in these foci. Interestingly, by exchanging UBDs between different proteins we have learned that each UBD seems to have a distinct functional role, surprisingly not limited to Ubbinding ability. In fact, swapping the UBZ of Wrnip1 with the UBM of polι abolished the localization of Wrnip1 to foci despite preserving the Ub-binding ability of the chimeric protein. In summary, this work provides an overview of how post-translation modification of proteins by Ub can regulate several DNA transactions. Firstly, key regulators (e.g. PCNA) can be differentially modified by Ub. Secondly, specialized UBDs (e.g. UBM, UBZ) embedded only in a subset of proteins act as modules able to recognize these modifications. Thirdly, by means of mediating auto-ubiquitylation, UBDs can modulate the behaviour of host proteins by allowing for either in cis or in trans Ub-UBD interactions.

This complex when applied to column chromatography, allow s the separation of various dehydrogenases as a result of their different complex stability coefficients. Alcohol dehydrogenase from liver, lactate dehydrogenase, and adenylate kinase, which all bind to the ADP-analog residues of the gel matrix, can thus be separated by different salt gradients. Alcohol dehydrogenase from yeast, however, does not form a complex and can easily be eluted from the column with phosphate buffer. Glyceraldehyde-3 phosphate and aldehyde dehydrogenases can be eluted by the addition of NAD+ or NADH to the buffer. The uncharged 1,4-dihydropyridin ring of the reduced coenzyme produces a more stable complex with the dehydrogenases than the oxidized form.
The title compound, C14H11NO4, crystallizes with two molecules in the asymmetric unit. The major conformational difference between these two molecules is the dihedral angle between the aromatic rings, namely 36.99 (5) and 55.04 (5)°. The nitro groups are coplanar with the phenyl rings to which they are attached, the O—N—C—C torsion angles being -1.9 (3) and 1.0 (3)° in the two molecules.
