Qualität und Qualitätsmanagement in der universitären naturwissenschaftlichen Lehrerfortbildung

Vor dem Hintergrund der politischen Entwicklungen in Hessen und der allgemeinen Debatte über Qualität im Bildungsbereich sollte im Lehrerfortbildungszentrum Chemie (lfbz-Chemie) der Universität Frankfurt am Main eine systematische Qualitätsentwicklung etabliert und dieses Vorhaben wissenschatflich begleitet werden. Die wissenschaftliche Arbeit besteht aus drei Säulen: - Eine erste, qualitative Studie sollte Qualitätskriterien und -indikatoren liefern. - In einer Fallstudie am lfbz-Chemie Frankfurt sollte die Einführung von Instrumenten des Qualitätsmanagements aus dem gewerblichen Weiterbildungsbereich beobachtet und bewertet werden. - Eine bundesweite Umfrage unter universitären Fortbildungsanbietern im naturwissenschaftlichen Bereich sollte sowohl die Struktur der anbietenden Institutionen als auch die allgemeine Einstellung der Befragten zum professionellen Qualitätsmanagement (QM) nach einem im gewerblichen Bereich geläufigen Modell (LQW) beleuchten. Die Qualitätsindikatoren und -kriterien fielen sehr spezifisch für die naturwissenschaftliche Lehrerfortbildung aus. Allein die drei folgenden Bereiche erfassen zwei Drittel aller Nennungen: die „Qualität der Teamer bzw. Moderatoren“, die „Fortbildungsgestaltung“ und die „Zielgruppenorientierung (Schulbezug)“. Nimmt man die Anzahl der Indikatoren und Kriterien pro Bereich als Maßstab, fokussieren die Befragten sehr stark auf Anforderungen, die die Professionalität des Personals betreffen. Zur Orientierung für die Qualitätsarbeit am lfbz-Chemie (Fallstudie) wurde das QM-Modell LQW 2 gewählt; als kleinere Instrumente kamen die Tabellen nach dem Vorbild der Balanced Scorecard (BSC) nach Kaplan und Norton sowie die Stärken-Schwächen-Analyse zum Einsatz. Als erstes Ergebnis der Fallstudie kristallisierten sich drei Arbeitsbereiche heraus: Personalorganisation, Evaluation und Innovationen. Diese Arbeitsfelder ergänzten die identifizierten Anforderungen an Fortbildung somit um Bereiche, die eher der organisatorischen Ebene zuzuordnen sind. Es ließ sich in der Fallstudie feststellen, dass ein ganzes Qualitätsmanagementmodell wie LQW 2 nur als Anregung für den einen oder anderen, als wichtig erachteten Bereich dienen konnte, während sich kleinere Instrumente eher als handhabbar erwiesen. Außerdem konnte postuliert werden, dass - ein persönlicheres Eingehen auf die Ziele und Aufgaben der einzelnen Personen im Sinne eines Total Quality Managements vermutlich zu besseren Ergebnissen geführt hätte als die Konzentration auf allgemeine Themen und die Orientierung am eher abstrakten Modell, - Erfolge sehr nachdrücklich kommuniziert werden müssen und - möglichst eine Person aus dem Stammpersonal für den Bereich Qualität verantwortlich sein sollte. Um ergänzend zur Fallstudie zu allgemeineren Schlüssen gelangen zu können, wurde eine Charakterisierung der universitären Fortbildungsanbieter mittels Clusteranalyse mit Daten der bundesweiten Umfrage durchgeführt. Sie zeigte vier Typen von Anbietern auf. Insgesamt fanden sich neben der Gruppe der großen Anbieter, zu denen auch das lfbz-Chemie gehört, nur vergleichsweise kleine Anbieter. Die in der Fallstudie erhaltenen Ergebnisse können somit nur eingeschränkt auf die meisten Anbieter übertragen werden. Insgesamt konnten Stärken und Schwächen der universitären Anbieter identifiziert werden, mit denen sich die universitäre naturwissenschaftliche Lehrerfortbildung von der gewerblichen Weiterbildung abhebt. Stärken lagen vor allem in den besonderen Kompetenzen, die an der Hochschule anzutreffen sind. Problemfelder lagen ausgerechnet hauptsächlich im Personalbereich. Als spezifische Schwächen der universitären Anbieter im Personalbereich kann z. B. die Stellung der Fortbildung als Nebenaufgabe oder der Verlust von Know-how durch die Fluktuation des Personals gelten. Auch allgemein kann ein Modell wie LQW 2 als Anregung für die Qualitätsarbeit in der universitären Lehrerfortbildung dienen, nicht jedoch umfassend umgesetzt werden. Die „kleineren“ Instrumente können dagegen eher als allgemein geeignet betrachtet werden. Während die „großen“ Anbieter durch verbesserte personelle Ressourcen für Aufgaben wie die Qualitätsarbeit unterstützt werden könnten, bietet sich für die „kleinen“ Anbieter eher die Unterstützung durch eine zentrale Stelle der Universität an, um die Fortbildungsanbieter doch noch in ein professionelles Qualitätsmanagement einbinden zu können.
Schwerpunkt der vorliegenden Arbeit war die Formulierungsentwicklung eines nanopartikulären Arzneistoffträgers für Zytostatika zur Gewinnung einer „Drug Targeting“ Zubereitung. Im Fokus der Arbeit stand dabei die Stabilität der unterschiedlichen Zubereitungen. Die untersuchten Partikel waren dabei auf Basis von humanem Serumalbumin. Mittels einer etablierten Desolvatationsmethode ließen sich reproduzierbar Nanopartikel im Größenbereich von 200 nm herstellen. Zur Partikelgrößenbestimmung bediente man sich sowohl der hotonenkorrelationsspektroskopie (PCS) als auch der Analytischen Ultrazentrifugation (AUZ). Bei der Untersuchung verschiedener HSA-Chargen, stellte sich heraus, dass das Ausgangsmaterial einen Einfluss auf die Partikelgröße besaß. Nichtsdestotrotz waren die Partikelgrößenschwankungen aufgrund von unterschiedlichen HSA-Chargen gering. Durch Einlagerung von Doxorubicin in die Partikelmatrix kam es zu einer wesentlichen Partikelvergrößerung, so dass die resultierten Partikel eine Größe von etwa 400 nm aufwiesen. Die Inkorporation des Arzneistoffs in die Partikelmatrix war reproduzierbar und stabil. Auch der Einsatz von Serumalbumin aus rekombinanter Quelle erwies sich als geeignet um Nanopartikel herstellen zu können. Allerdings waren unbeladene Partikel aus diesem Material wesentlich größer als Nanopartikel, die aus dem Standardmaterial hergestellt wurden. Doxorubicinbeladene Partikel zeigten wiederum eine vergleichbare Größe wie Partikel aus HSA. Allerdings war die Beladung der rHSA-Nanopartikel geringer als die der SA-Nanopartikel. Dies zeigte sich besonders bei der geringeren Quervernetzung von 40%. Als Fazit lässt sich sagen, dass prinzipiell eine Herstellung von sowohl Leerpartikeln als auch Doxorubicin-beladenen Partikeln möglich war. Die Biodegradierbarkeit von Nanopartikeln ist eine wichtige Voraussetzung für einen therapeutischen Einsatz kleinpartikulärer Strukturen, damit diese nicht im Körper kumulieren. Zudem könnte bei nicht abbaubaren Partikeln die Idee der intrazellulären Freigabe des eingebetteten Arzneistoffs aus der Partikelmatrix nicht verwirklicht werden. Vorversuche haben gezeigt, dass sich Nanopartikel auf Basis von HSA mit einer Reihe von Enzymen abbauen lassen. Versuche, Nanopartikel aus rHSA enzymatisch abzubauen, führten zu vergleichbaren Abbaukinetiken wie bei Partikeln aus HSA. Zu den eingesetzten nzymen zählten Proteinase K, Protease, Trypsin, Pankreatin, Pepsin und Cathepsin B, die mit Partikel aus rHSA mit den Quervernetzungen 40, 60, 80 und 100% inkubiert wurden. Alle Abbaukinetiken zeigten dabei, dass die Abbaugeschwindigkeit vom Grad der Quervernetzung abhängig war. 40% quervernetzte Nanopartikel wurden am schnellsten, 100% quervernetzte Partikel am langsamsten enzymatisch degradiert. Die Abbauversuche mit Cathepsin B bei zwei verschiedenen pH-Werten zeigten, dass die Wahl des richtigen pH-Werts entscheidend für einen effektiven Abbau ist, denn nur bei einem pH-Wert von 5,4 wurden die Nanopartikel von Cathepsin B abgebaut. Im Gegensatz dazu, wurden die Nanopartikel bei pH 6,4 kaum degradiert. Des Weiteren wurden Beladungsversuche von HSA-Nanopartikeln mit Cisplatin durchgeführt. Dabei wurden im ersten Schritt Adsorptionsversuche an gelöstes HSA durchgeführt, da viele Arzneistoffe eine hohe Plasmaeiweißbindung besitzen, wenn sie sich im Blutkreislauf des Menschen befinden. Diese Tatsache sollte bei der Herstellung von arzneistoffhaltigen Nanopartikel auf Basis von humanem Serumalbumin ausgenutzt werden. Vor dem eigentlichen Desolvatationsprozess wurden gelöstes HSA und Cisplatin bei unterschiedlichen pH-Werten und für unterschiedliche Zeitintervalle inkubiert, um eine Adsorption des Cisplatins an das Protein zu erreichen. Dadurch soll es bei der anschließenden Desolvatation zu einer effektiveren Inkorporation des Arzneistoffs kommen. Die Ergebnisse haben gezeigt, dass bei sauren pH-Werten die Adsorption schwächer ausfällt als bei einem pH-Wert von 8,0. Zudem war die Adsorption umso ausgeprägter, je länger die Inkubation stattfand. Bei einem pH-Wert von 8,0 führten steigende Konzentrationen an eingesetztem Cisplatin bei konstanter Menge an HSA, zu höheren Konzentrationen an adsorbiertem Arzneistoff. Prozentual gesehen, führten aber zunehmende Mengen an eingesetztem Cisplatin zu geringeren Adsorptionsraten. Als Fazit muss aber festgehalten werden, dass Cisplatin eine geringe Tendenz zur Adsorption an gelöstes HSA zeigte. Die Herstellung von Cisplatin-beladenen HSA-Nanopartikeln zeigte, dass die Desolvatation bei pH 8,0 zu guten Ergebnissen führte. Zum einen wiesen die erhaltenen Nanopartikel gute physikochemische Eigenschaften mit nahezu quantitativen Partikelausbeuten auf, zum anderen besaßen die Partikel eine hohe Beladungseffizienz. Dabei galt, je höher die eingesetzte Menge an Cisplatin war, umso mehr Cisplatin wurde in die Matrix der Partikel eingelagert. Die Dauer der zuvor stattfindenden Adsorption spielte dabei eine eher untergeordnete Rolle im Vergleich zu den Adsorptionsversuchen. Eine Erklärung für diese Beobachtung könnte sein, dass die Zugabe des Desolvatationsmittels Ethanol, in welchem Cisplatin sehr schwer löslich ist, zu einer verstärkten Interaktion zwischen Arzneistoff und Protein führt und diese Komponenten zusammen ausfallen, nahezu unabhängig von der zuvor stattfindenden Adsorptionsphase. Um die Idee einer „Drug Targeting“ Zubereitung umsetzen zu können, wurden mit Hilfe von Polyethylenglykol-Ketten sowohl Leerpartikel als auch arzneistoffbeladene Nanopartikel auf ihrer Oberfläche mit monoklonalen Antikörpern modifiziert. Als Verum wurde dabei Trastuzumab verwendet, als Kontrolle diente ein IgG-Antikörper von Sigma, der kein Target besitzt. Sowohl eine adsorptive Bindung als auch die kovalente Kopplung der Antikörper an die Oberfläche der nanopartikulären Strukturen konnte reproduzierbar durchgeführt werden. Dabei spielte es keine Rolle, ob die Partikel mit Arzneistoff beladen waren oder nicht. Trastuzumab zeigte ein hohes Maß an adsorptiver Bindung an die Oberfläche von HSA-Nanopartikeln, die bei dem Kontollantikörper nicht festgestellt werden konnte. Von allen Partikelpräparationen wurden die Partikelgröße, die Größenverteilung, das Zetapotential und die Partikelausbeute bestimmt. Durch das Aufbringen neuer Oberflächenstrukturen, kam es zu keiner wesentlichen Veränderung der Partikelgröße bzw. der Oberflächenladung. Durch die zahlreichen Umsetzungsschritte, die für eine Oberflächenmodifikation nötig sind, kam es bei Nanopartikeln ohne Arzneistoffbeladung zu einem Verlust an Partikelausbeute. Da die Doxorubicin-beladenen Partikel viel größer waren als Leerpartikel, ließen sie sich einfacher und effektiver abzentrifugieren, was in einem geringeren Partikelausbeuteverlust sichbar wurde. Da die Gefriertrocknung zu den Standardmethoden zählt Zubereitungen eine gute Haltbarkeit zu verleihen, wurden eine Reihe von nanopartikulären Zubereitungen der Lyophilisation unterzogen und im Hinblick auf ihre Langzeitstabilität unter verschiedenen Lagerungsbedingungen getestet. Dabei stellte sich heraus, dass es mittels Gefriertrocknung möglich war aus HSA-Nanopartikelsuspensionen einfach und reproduzierbar Lyophilisate herzustellen. Als geeignete Hilfsstoffe kristallisierten sich dabei die Zucker Sucrose, Trehalose, der Zuckeralkohol Mannitol und Emulgatoren wie Tween® 80 und Pluronic® F68 heraus. Als ungeeignet zeichnete sich der Einsatz von L-Arginin und eines Natriumphosphat-Puffers pH 8,0 ab. Larginin führte schon vor dem Gefriertrocknen zu einer Partikelvergrößerung, die nach der Lyophilisation noch ausgeprägter war. Puffer auf Basis von Natriumsalzen führen zu einem starken pH-Shift während des Gefriertrocknungsprozesses. Dies führte beim Einsatz des Phosphat-Puffers pH 8,0 zu einem starken Partikelwachstum. Gefriertrocknungsprozesse mit unterschiedlichen Geräten haben gezeigt, dass der Zusatz von Sucrose bzw. Trehalose oder Mannitol ab einer Konzentration von 2% (m/V) zu guten physikochemischen Eigenschaften der rekonstituierten Proben führte. Hilfsstoffkombinationen, wie sie in der Literatur beschrieben sind, waren für die Stabilisierung der nanopartikulären Strukturen nicht nötig. Die Langzeitlagerungsstabilitätsdaten der gefriergetrockneten HSA-Nanopartikel über 13 Wochen bei unterschiedlichen Temperatur- und Luftfeuchtigkeitsbedingungen zeigten eine Überlegenheit der Hilfsstoffe Sucrose und Trehalose im Vergleich zu Mannitol. Als geeignete Hilfsstoffkonzentration stellte sich hier ein 3%iger (m/V) Zusatz heraus. Versuchsansätze mit unterschiedlichen Gefriertrocknern und somit unterschiedlichen Prozessen zeigten, dass nicht nur die Auswahl der Hilfsstoffe, sondern auch die Bedingungen des Gefriertrocknungsprozesses Einfluss auf die Lagerungsstabilität hatten. Ein kontrollierter Prozess zeigte sich dabei gegenüber dem schnellen Einfrieren mittels Stickstoff als überlegen. Von Nanopartikelsuspensionen mit den Hilfsstoffen Trehalsoe, Sucrose und Mannitol wurden die Glasübergangstemperaturen bestimmt. Die Ergebnisse deckten sich im Wesentlichen mit den in der Literatur beschriebenen Daten, so dass schlussgefolgert werden kann, dass die HSA-Nanopartikel keinen wesentlichen Einfluß auf die Glasübergangstemperatrur hatten. Zusätzlich wurde die Restfeuchte der Lyophilisate direkt nach der Gefriertrocknung und nach 13 wöchiger Einlagerungszeit bestimmt. Die Proben wiesen direkt nach dem Prozess eine Restfeuchte von ungefähr 3% auf, durch Lagerung der Partikel bei erhöhter Luftfeuchtigkeit kam es zu einem Anstieg des Wassergehalts in den Proben. Mit den Hilfsstoffzusätzen Trehalose, Sucrose und Mannitol ließen sich auch Doxorubicin-beladene HSA-Nanopartikel gefriertrocknen. Dabei kam es nach der Rekonstitution dieser Partikel zu keinem Austreten des eingelagerten Arzneistoffs. Zusätzlich wurden oberflächenmodifizierte Partikel lyophilisiert. Dabei bestand die Modifikation zum einen aus Methoxypolyethylenglykol-Ketten. Zum anderen wurden über NHS-PEG-Mal Crosslinker kovalent monoklonale Antikörper auf die Oberfläche von HSANanopartikeln gebunden. Zusätzlich wurden auch HSA-NP in Suspension einer Untersuchung bezüglich Langzeitlagerungsstabilität unterworfen. Dabei wiesen auch HSA-Nanopartikel in Suspension bei verschiedenen Einlagerungsbedingungen eine hohe Stabilität auf. Partikel, die über einen Zeitraum von 210 Tagen eingelagert wurden, zeigten bei den Temperaturen 4°C, 20°C und 30°C im Hinblick auf Partikelgröße und Polydispersität kaum Veränderungen. Die Lagerung der Nanopartikel bei Minusgraden führte allerdings zu Mikropartikeln. Bei der Untersuchung der Partikelüberstände hinsichtlich herausgelöstem HSA zeigte sich, dass je höher die Lagerungstemperatur und je länger die Einlagerung war, umso mehr HSA löste sich aus der mittels Glutaraldehyd fixierten Matrix heraus. Bei den eingefrorenen Partikeln löste sich über die gesamte Lagerungszeit kein Protein aus den Nanopartikeln heraus. Zellkulturexperimente zeigten, dass im Gegensatz zu Kontrollzubereitungen, Nanopartikel, die an ihrer Oberfläche therapeutisch wirksame Antikörper trugen, spezifisch von Krebszellen aufgenommen wurden und im Zellinneren den eingebetteten Arzneistoff freisetzten. Daraus resultierte eine spezifische Toxizität dieser Zubereitungen gegenüber Tumorzellen. Dies ist ein erster Ansatz, um zeigen zu können, dass durch nanopartikuläre Trägersysteme die unerwünschten Nebenwirkungen der unspezifisch wirkenden Zytostatika reduziert werden können. In weiteren Versuchen, vor allem mit Hilfe von in vivo Versuchen muss gezeigt werden, dass das Partikelsystem stabil genug ist, ausreichend lang im Körper zirkulieren zu können. Dies ist nur möglich, wenn die Nanopartikel in der Lage sind, das Gefäßsystem im Bereich des Tumorgewebes zu verlassen und anschließend in die Tumormasse einwandern können. Nur dann können die in den Zellkulturversuchen gezeigten Effekte greifen.
The synthesis of [Ph4As+]2[Cl4Re(NS)(NSCl)2-] · CH2Cl2 (4) from the reaction of S4N4, Cl4ReN, and Ph4AsCl is reported. CH2Cl2 is used as solvent. The reaction of S4N4 with Re2Cl10 similarly leads to the salt [Ph4As+][Cl2ReNS-] (5) in a smaller yield. 4 crystallizes in the triclinic space group P1̅ with Z = 2, a - 10.434(2), b = 12.1454(6), c = 21.125(2) Å, a = 81.210(6), β = 86.70(1), γ = 76.624(8)°.
In eukaryotic cells, mitochondria host ancient essential bioenergetic and biosynthetic pathways. LYR (leucine/tyrosine/arginine) motif proteins (LYRMs) of the Complex1_LYR-like superfamily interact with protein complexes of bacterial origin. Many LYR proteins function as extra subunits (LYRM3 and LYRM6) or novel assembly factors (LYRM7, LYRM8, ACN9 and FMC1) of the oxidative phosphorylation (OXPHOS) core complexes. Structural insights into complex I accessory subunits LYRM6 and LYRM3 have been provided by analyses of EM and X-ray structures of complex I from bovine and the yeast Yarrowia lipolytica, respectively. Combined structural and biochemical studies revealed that LYRM6 resides at the matrix arm close to the ubiquinone reduction site. For LYRM3, a position at the distal proton-pumping membrane arm facing the matrix space is suggested. Both LYRMs are supposed to anchor an acyl-carrier protein (ACPM) independently to complex I. The function of this duplicated protein interaction of ACPM with respiratory complex I is still unknown. Analysis of protein-protein interaction screens, genetic analyses and predicted multi-domain LYRMs offer further clues on an interaction network and adaptor-like function of LYR proteins in mitochondria.
Cytochrome c oxidase (CcO), also called Complex IV of the aerobic respiratory chain, is located in the plasma membrane of prokaryotes and in the inner mitochondrial membrane of eukaryotes. The redox energy of dioxygen reduction is used to translocate protons across the membrane resulting in an electrochemical proton gradient. The generated proton gradient is exploited by the adenosine-5’-triphosphate synthase. In this work, bacterial four-subunit aa3-Type CcO from Paracoccus denitrificans (ATCC 13543, 4 SU-wt ATCC CcO) was used for analyses. 1) The recombinant homologously produced 4 SU-wt CcO (4 SU-wt rec CcO) was functionally compared with the native 4 SU-wt ATCC CcO. The 4 SU-wt rec CcO showed functional deficiencies as determined by UV-vis spectroscopy and electron paramagnetic resonance (EPR) studies. Total X-ray Reflection Fluorescence measurements show in both wild type CcOs the same ratio of the redoxactive Fe and Cu (2 Fe : 3 Cu) indicating full complement of the functional metals. If CcO contains only subunit I and II, it loses its functional integrity during continuous turnover activity. The importance of subunit III for integrity of CcO was demonstrated using 2 SU-wt rec CcO. Crystallisation trials of suicide inactivated 2 SU-wt rec CcOs have been ineffective using standard crystallisation conditions. Crystals of active 2 SU-wt rec CcO (positive control) have been obtained under these conditions and this result indicates possible structural changes in suicide inactivated 2 SU-wt rec CcO. The structure of active 2 SU-wt rec CcO was determined to 2.25 Å resolution. 2) Terminal oxidases require four electrons for the cleavage of the dioxygen bond (O=O). In general, the catalytic cycle of CcO is described by the electron input and thus by the different redox states of the metal centres: the O, E, R, P and F state. The two-electron reduced R intermediate is able to donate four electrons for dioxygen reduction forming the P state. The P intermediate is an oxoferryl state implying the lack of an electron for the R -> P transition, because the metal centres can only provide three electrons (Fe+II forms Fe+IV and Cu+II forms Cu+I). The P state, where the dioxygen bond is already broken, shows an oxoferryl state (FeIV=O2-) and a nearby tyrosine is proposed to form a tyrosyl radical representing the donor of the missing electron. H2O2-induced artificial intermediates provide the opportunity to investigated different catalytic intermediates in detail. Mixing equimolar amounts of H2O2 to CcO in the O state induces the "two-electron" reduced PH state at high pH and the electronically equal "two-electron" reduced F• H state at low pH. The addition of an excess amount of H2O2 leads to the three-electron reduced FH state. Functional studies using the 4 SU-wt ATCC CcO have demonstrated a bound peroxide (O- - O-) intermediate during the catalytic cycle. Using EPR it was previously shown that Y167 hosts a radical species in PH/F• H state which suggests that Y167 could provide this "missing electron". While X-ray structural models of CcO and Fourier-transformed infrared (FTIR) measurements of oxygenated ("pulsed") 4 SU-wt ATCC CcO suggest a bound peroxide in the O state, UV-vis and EPR spectroscopic studies indicate that other intermediates may also contain such peroxide species. Equimolar and excess amounts of H2O2 induce the PH/F• H and FH states, respectively and catalase treatment of the FH state leads, contrary to the natural direction of the catalytic cycle, to the apparent transition of the FH -> PH/F• H states, which is accompanied by reappearance of an EPR signal from the Y167• radical. The novel PFH/F• FH states are presented here and we postulate that the FH state hosts a superoxide (or peroxide) adduct at CuB in the binuclear site. In addition, the novel P10 state is also introduced having a maximum at lambda = 612 nm in the difference absorption spectrum (minus the O state). The P10 state is induced by mixing CcO in the O state with a pH 10 buffer. This pH 10 induced state resembles standard P states such as PCO, PH and PR. However, the P10 state evolves out of the O state without addition of reduction equivalents. Using EPR spectroscopy it was shown that Y167 hosts a radical species in the P10 state such as in the PH state. In summary, all functional data presented here provide evidence for a peroxide bound during the O state. Finally, a new model for the natural catalytic cycle is proposed. If the O state contains a peroxide, it is also likely that the E and R state contain this species. Even the oxoferryl intermediates P and F states may complex a peroxide at CuB in the binuclear site. 3) The amino acid residue Y167, which hosts the radical in the PH/F•H states, is not directly part of the binuclear site of CcO. For identification of the primary electron donor, two tryptophan variants of CcO, W272F and W164F, which are located nearby the binuclear site, were produced. Evidence is provided that W272 is a kinetically fast electron donor for the O2 molecule. The electron is replenished by Y167, or probably by Y280 in the natural cycle. The Y167 radical is detectable by EPR spectroscopy after treatment with equimolar amounts of H2O2 in the active variant W164F, but is absent in the inactive variant W272F. 4) CcO contains two proton conducting pathways, the D- and the K-pathway. Proteoliposomes of the variants H28A and D30N, mutations located at the entrance of the D-pathway, both show the identical proton pumping activity as the 4 SU-wt rec CcO (pumped H+/e- = 1). The variant N113D shows abolished proton pumping (pumped H+/e- = 0), but a relative high cytochrome c oxidation activity (63 %). G196D displays no cytochrome c oxidation and proton pumping activity. Overall, the addition or removal of a negative charge within the D-pathway such as in D124N, N131D, N113D and G196D leads to a decoupled phenotype indicating the high degree of electrostatic coupling in CcO.
A simple and fast method of lipid analysis of isolated intact mitochondria by means of MALDI-TOF mass spectrometry is described. Mitochondria isolated from bovine heart and yeast have been employed to set up and validate the new method of lipid analysis. The mitochondrial suspension is directly applied over the target and, after drying, covered by a thin layer of the 9-aminoacridine matrix solution. The lipid profiles acquired with this procedure contain all peaks previously obtained by analyzing the lipid extracts of isolated mitochondria by TLC and/or mass spectrometry. The novel procedure allows the quick, simple, precise, and accurate analysis of membrane lipids, utilizing only a tiny amount of isolated organelle; it has also been tested with intact membranes of the bacterium Paracoccus denitrificans for its evolutionary link to present-day mitochondria. The method is of general validity for the lipid analysis of other cell fractions and isolated organelles.
Strukturelle und funktionelle Untersuchungen am Cytochrom-bc1-Komplex aus Paracoccus denitrificans

Cytochrom bc-Komplexe sind zentrale Enzyme energietransduzierender Elektronen-transportketten. Anerkanntes Funktionsprinzip ist der Q-Zyklus; mechanistische Details insbesondere der Chinoloxidation am Qo-Zentrum sind noch unklar. Ein Verständnis des Qo-Zentrums ist auch von Interesse, da hier Inhibitoren in Form von Fungiziden und Malariatherapeutika wichtige Anwendung finden. In den vergangenen Jahren wurde eine Reihe mitochondrialer Komplexe kristallographisch charakterisiert, die Struktur eines bakteriellen Enzyms steht jedoch aus. Hauptziel dieser Arbeit war es, Ansätze zur Strukturaufklärung des bc1-Komplexes aus Paracoccus denitrificans (P.d.) zu finden, der als homologes Enzym mitochondrialer Komplexe bei einfacher genetischer Zugänglichkeit ein wichtiges Modellsystem darstellt. In der vorliegenden Arbeit wurde auf die Kristallisation des bc1-Komplexes aus S. cerevisiae aufgebaut, die mit Hilfe monoklonaler Antikörperfragmente (Fv) gegen die Rieske-Untereinheit (ISP) erreicht wurde; die Fv-Fragmente erleichtern die Ausbildung von Kristallkontakten. Die Epitopregion des Hefeenzyms wurde genetisch auf den bakteriellen Komplex übertragen, um dessen Ko-Kristallisation mit dem bereits verfügbaren Fv zu ermöglichen. Punktuelle Anpassungen führten zu keiner signifikanten Bindung, ein weitergehender Austausch des entsprechenden ISP-Bereichs verbesserte die Bindung hingegen deutlich. Die besten Ergebnisse konnten mit chimären Enzymen erzielt werden, bei denen die gesamte ISP-Ektodomäne durch das Hefe-Homologe ersetzt wurde. Aufbauend auf dieser Arbeit scheint die Fv-vermittelte Kristallisation des Enzyms ein greifbares Ziel. Eine komplementäre Strategie zielte auf die strukturelle Charakterisierung der Rieske-Ektodomäne (ISF). Das klonierte ISF wurde in E. coli überexprimiert, lag jedoch fast vollständig in inclusion bodies vor. Das ISF konnte in eine lösliche Form rückgefaltet werden, die anschließende chemische Rekonstitution zum Holo-Protein gelang jedoch nur mit einer Ausbeute von ~ 1 %. Die geringe Menge an löslichem ISF, die sich nach Expression in E. coli isolieren lässt, trägt kein [2Fe-2S]-Zentrum. Durch Koexpression der für die Biogenese von Eisen-Schwefel-Zentren relevanten Gencluster konnte die lösliche ISF-Fraktion in vivo in die Holo-Form konvertiert werden. Auch hier war aber die Gesamtausbeute für strukturelle Untersuchungen zu gering. Eine homologe Expression in P.d. war nur für das komplette ISP nachweisbar, nicht für das verkürzte ISF. Durch gerichtete Mutagenese konnte hier erstmals gezeigt werden, dass das bakterielle Rieske-Protein über den Tat-Translokationsweg in die Membran inseriert. Die Kristallstrukturen mitochondrialer bc1-Komplexe zeigen ein dimeres Enzym. Die Assoziation des bakteriellen Komplexes wurde in dieser Arbeit mit der analytischen Ultrazentrifugation untersucht, und auch hier wurde eindeutig ein Dimer nachgewiesen. Dynamische Messverfahren deuteten jedoch auf einen höheren Assoziationszustand hin. Es bleibt unklar, ob diese Diskrepanz durch Formparameter oder die Detergenzbindung begründet ist oder ob unter bestimmten Versuchsbedingungen möglicherweise Tetramere vorliegen. In situ ist der bc1-Komplex strukturell mit den Komplexen I und IV sowie dem Elektronenüberträger Cyt c552 assoziiert. Die Analyse verschiedener Deletionsstämme zeigte, dass Komplex I der Atmungskette durch diesen Superkomplex stabilisiert wird. Dieser Befund konnte kürzlich in anderen Arbeiten auch an menschlichen Mitochondrien bestätigt werden. Neben strukturellen Aspekten wurden am bc1-Komplex auch die H+-Translokation und die Chinonbindung untersucht. Da chemische Modifikationsexperimente zeigen, dass ein saurer Rest im ISP eine kritische Rolle für die Kopplung von H+-Translokation und Elektronentransport spielt, wurden durch gerichtete Mutagenese kombinatorisch saure Reste gegen entsprechende Säureamide ersetzt. Die biochemische Charakterisierung wurde nur für eine Fünffach-Mutante durchgeführt; diese erwies sich jedoch als zu instabil, um verlässliche Daten aus H+-Pumpexperimenten zu gewinnen. Die Charakterisierung der übrigen Mutanten scheint lohnenswert, da der relevante Aminosäurerest auch in anderen Arbeiten noch nicht identifiziert werden konnte. Der Gehalt des aufgereinigten Enzyms an spezifisch gebundenem Chinon wurde FTIR-spektroskopisch quantifiziert. Eine Stöchiometrie von ~ 3 Chinonmolekülen/Monomer stützt das double occupancy-Modell, demzufolge zwei Substratmoleküle am Qo-Zentrum binden; das dritte Chinon bindet am Qi-Zentrum. Partielle Extraktion des Chinons und Messungen bei verschiedenen pH-Werten zeigten, dass die Substratbindung mit Protonierung eines sauren Rests einhergeht. Möglicherweise handelt es sich dabei um Glu295 des Cytochrom b, das auf Basis der Kristallstrukturen als primärer H+-Akzeptor am Qo-Zentrum diskutiert wird. Aufbauend auf dieser Arbeit können die an der Chinonbindung beteiligten Reste durch Mutagenese identifiziert werden.
MHC Klasse I Moleküle liegen im Endoplasmatischen Reticulum (ER) als Dimer bestehend aus einer schweren Kette mit Transmembrandomäne und einem 12 kDa-Protein, dem ß2-Mikroglobulin vor. Nach der Beladung des MHC-Klasse I-Moleküls mit einem antigenen Peptid, welche vorwiegend im Cytosol durch proteasomalen Abbau generiert und durch den Transportkomplex TAP ins ER transloziert werden, findet der Transport des MHC-Peptid-Komplexes zur Zelloberfläche statt. Dort wird das Antigen cytotoxischen T-Zellen präsentiert. An der Assemblierung und Reifung von MHC-Klasse I-Molekülen sind verschiedene Chaperone beteiligt. Eine wichtige Rolle beim Peptidbeladungsprozess von MHC-Klasse I-Molekülen spielt Tapasin. Dabei handelt es sich um ein 48 kDa, MHC-codiertes Typ I Transmembran-Glycoprotein aus der Immunglobulinsuperfamilie. Es verbrückt den TAP-Komplex mit MHC-Klasse I-Molekülen, hält unbeladene MHC-Klasse I-Moleküle im ER zurück und führt eine Qualitätskontrolle des gebundenen Peptids durch. Bei dieser Peptideditierung werden Peptide wieder selektiv aus der MHC-Bindungstasche entfernt, wenn sie mit einer niedrigen Affinität gebunden sind. Dadurch wird sichergestellt, dass keine leeren oder suboptimal beladenen MHC-Peptid-Komplexe an die Zelloberfläche gelangen. In der vorliegenden Arbeit wurde ein System etabliert, mit dem der Einfluss von Tapasin auf die Peptidbeladung von MHC-Klasse I-Molekülen in vitro untersucht werden kann. Dazu wurde ein Verfahren zur heterologen Expression und Reinigung von funktionalem, löslichem Tapasin aus E. coli-Zellen aufgestellt. Weiterhin wurden ß2m und die schwere Kette von HLA-B*2705 heterolog in E. coli-Zellen exprimiert, isoliert und zusammen mit einem Reporterpeptid zum funktionalen HLA-B*2705 renaturiert. Bei Untersuchungen der Wechselwirkung zwischen Tapasin und HLA-B*2705-Molekülen konnte mittels der Oberflächen-Plasmonen-Resonanz-Spektroskopie eine direkte Interaktion zwischen Tapasin und unbeladenem HLA-B*2705 gezeigt werden. Detailliertere Untersuchungen zur Rolle von Tapasin bei der Peptidbeladung wurden mittels einer Gelfiltration gekoppelt mit der Fluoreszenzdetektion des Reporterpeptids durchgeführt. Dabei konnte festgestellt werden, dass unbeladene MHC-Klasse I-Moleküle in Anwesenheit von Tapasin stabilisiert werden und in einer Konformation gehalten werden, die eine Assoziation mit Peptid fördert. Weiterhin wurde gezeigt, dass durch Tapasin die Assoziationsrate für die Peptidbindung erhöht ist. Somit kann in Anwesenheit von Tapasin eine größere Anzahl an Peptiden auf eine stabile und hochaffine Bindung an MHC-Klasse I-Moleküle überprüft werden. In Experimenten mit bereits beladenen MHC-Klasse I-Molekülen konnte gezeigt werden, dass der neugebildetete Komplex auch nach erfolgter Peptidassoziation durch Tapasin stabilisiert wird. Dies ist vermutlich auf eine Erniedrigung der Dissoziationsrate für das Peptid zurückzuführen. Mit dem in dieser Arbeit etablierten Untersuchungssystem ist die Grundlage zu detaillierten Studien der Rolle von Tapasin bei der Peptidbeladung von MHC-Klasse I-Molekülen geschaffen.
Im Rahmen dieser Arbeit werden die Synthese, Eigenschaften und Anwendungsmöglichkeiten von Arylalkyl-Rückgrat modifizierten DNA-Oligonucleotiden untersucht. Das erste Ziel der vorliegenden Arbeit war, lipophile, arylalkylmodifizierte Oligonucleotide zu synthetisieren und die Auswirkungen der absoluten Konfiguration der Modifikationen auf die Eigenschaften der resultierenden Duplexe zu untersuchen. Als zweites sollten die Modifikationen in Antisense-Oligonucleotide eingebaut werden um diese auf ihre Anwendbarkeit für die lnhibierung der HCV Genexpression zu testen. Im Rahmen dieser Arbeit wurden 18 unterschiedliche Rückgrat-Modifikationen synthetisiert. Dabei wurde die Alkylkettenlänge wie auch die Größe des aromatischen Systems variiert. Zudem wurde untersucht, welchen Einfluss Ringsubstituenten auf die Eigenschaften der resultierenden Oligonucleotide ausüben. Die Rückgratmodifikationen wurden über die Festphasensynthese nach der Phosphoramiditmethode in Oligonucleotide eingebaut. Als Ausgangsverbindungen für die modifizierten Phosphoramidite dienten die Arylalkylhalogide. Diese wurden in einer dreistufigen in situ Reaktion - über das Grignard-Reagenz zu der entsprechenden cadmiumorganischen Verbindung und deren weitere Reaktion mit Phosphortrichlorid - zu den Arylalkyldichlorphosphanen umgesetzt. Die als Phosphorylierungsreagenzien fungierenden (Arylalkyl)(diisopropylamin)-chlorphosphane konnten durch Umsetzung mit N,N-Diisopropylamin erhalten werden. Die folgende Reaktion mit den 5'-hydroxyl- und aminogeschützten, natürlichen Nucleosiden führte zu den modifizierten Phosphoramidit-Bausteinen. Diese wurden mittels der OligonucleotidFestphasensynthese selektiv, an verschiedenen Positionen in sehr guten Ausbeuten in ModellOligonucleotide eingebaut und die erhaltenen Diastereoisomeren mittels RP-HPLC getrennt. Die einfach modifizierten, diastereoisomerenreinen Oligonucleotide zeigten eine signifikant erhöhte Lipophilie im Vergleich zu den unmodifizierten Strängen. Die Lipophilie nahm bei der Verlängerung der Alkylkettenlänge und der Vergrößerung des aromatischen Ringsystems pro (CH2)-Gruppe sowie pro weiterem Sechsring in konstanten Schritten zu, wodurch die Lipophilie gezielt gesteuert werden kann. Um den Einfluss der Modifikationen im Doppelstrang zu untersuchen wurden die Tm-Werte der Duplexe bestimmt und diese zudem CD- und Fluoreszenzspektroskopisch untersucht. Die erhaltenen Tm-Werte variierten sehr stark in Abhängigkeit der Alkylkettenlänge, der Ringgröße und der absoluten Konfiguration. Mit den Rp-konfigurierten benzyl- (B), (naphth-1-yl)methyl- (I) und 2,4-difluorbenzylmodifizierten (M) Oligonucleotid-Duplexen konnte eine Schmelzpunktserhöhung erzielt werden. Auch konnte mit den 3-(Anthracen-9-yl)propylphosphonaten K eine signifikante Tm-Wert Steigerung aufgrund eines "Dangling-End-Effektes" beobachtet werden. Die erhaltenen Tm-Werte korrelierten hervorragend mit den erhaltenen CD- und Fluoreszenz-Daten. Für die Zuordnung der absoluten Konfiguration der Modifikation wurden drei 3-Phenylpropylphosphonat-Dimere E synthetisiert. Die Zuordnung erfolgte mittels der 2D-ROESY-NMR-Spektren und den berechneten Protonenabständen der diastereoisomerenreinen Dimere sowie über empirische Regeln die von den Methylphosphonaten S abgeleitet wurden. Diese Ergebnisse lassen sich auf längere Oligonucleotide übertragen. Neben den Untersuchungen der Charakteristika der Arylalkyl-Rückgrat modifizierten Oligonucleotide wurden während dieser Arbeit einige Modifikationen gezielt auf ihre Einsetzbarkeit für den Antisense-Einsatz getestet. Als RNA-Zielsequenz wurden die Nucleotide 326-342 der 5'-nicht codierenden Region des Hepatitis C Virus gewählt. Im Rahmen dieser Arbeit wurden fünf unterschiedlich modifizierte Antisense-Oligonucleotide synthetisiert. Die arylalkylmodifizierten Oligonucleotide zeigten gute Hybridisierungseigenschaften gegenüber der sense-DNA bzw. sense-RNA und eine deutlich erhöhte Stabilität gegenüber der Nuclease Pl. Ferner konnte die Lipophilie der Oligonucleotide signifikant gesteigert werden. Die 2-Phenylethylphosphonate (D) und 2,4-Difluorbenzylphosphonate (M) sind zudem in der Lage die RNase H zu aktivieren. Alle dargestellten Antisense-Oligonucleotide wurden in einem zellfreien in vitro- sowie in einem in vitro-Zellkultur-Translations-Assay auf ihr lnhibierungspotential gegen die Hepatitis C Virus Genexpression getestet. Dabei zeigten die Benzylphosphonate (B), Phosphorthioate (Ps) und die 2-Phenylethylphosphonate (D) im zellfreien in vitro Testsystem hohe, spezifische Inhibierungsraten (>87%), bei einer Oligonucleotid-Konzentration von 5 µM. Auch erwiesen sich die arylalkylmodifizierten Antisense-Oligonucleotide, mit Ausnahme der 4-Phenylbutylphosphonate F, als sehr gute lnhibitoren der HCV-Genexpression in CCI13- und HepG2-Zellen.
