Gegenstand der hier vorgestellten Arbeit ist eine Applikation für die virtuelle Realität (VR), die in der Lage ist, die Struktur eines beliebigen Textes als begehbare, interaktive Stadt zu visualisieren. Darüber hinaus bietet das Programm eine besondere Textsuche an, die so in anderen konventionellen Textverarbeitungsprogrammen nicht vorzufinden ist. Dank der strukturellen Analyse und der Verwendung einiger außergewöhnlicher Analysetools des TextImager [2], ermöglicht text2City nicht nur die Suche nach bestimmten Textmustern, sondern zum Beispiel auch die Bestimmung der Textebene (Wort, Satz, Absatz, etc.) und einiges mehr. Ein weiteres Feature ist die Kommunikationsverbindung zwischen dem TextAnnotator-Service [1] und text2City, die dem Benutzer die Möglichkeit zum Annotieren bietet, aber auch von anderen Personen durchgeführte Annotationen sofort sichtbar machen kann. Für die Ausführung des Programms ist eine der beiden VRBrillen, Oculus Rift oder HTC Vive, ein für VR geeigneter PC, sowie die Software Unity nötig.
Between 1996 and 2006 the vegetation succession in drift sands and in blown-out gravel-rich depressions, located in the nature reserve Hulshorsterzand in the central Netherlands, was studied. Within this Natura 2000 habitat (type 2330: inland dunes with open Corynephorus and Agrostis grasslands) so-called lichen steppes are included, famous for their biodiversity, both in flora (cryptogams), and in fauna. With multivariate analysis, the relation between the primary succession, the species composition of the vegetation and the soil quality was studied in three different biotopes i.e. 1. drift sand, 2. blownout gravel-rich depressions and 3. drift sand after management. With a total of up to 34 species, lichen diversity in the study area appeared to be high. Biotope 1, i.e. the pioneer vegetation with Corynephorus canescens on blowing sand, is rather scarce in lichens. Between 1996 and 2006 green algae, C. canescens and Polytrichum piliferum appeared, with the result that much of the former open drift sand area was covered by pioneer stages of the Spergulo-Corynephoretum. If sand stopped blowing in, a combination of P. piliferum with the neophyte Campylopus introflexus occurred. The terminal stages of lichen succession on drift sand, a vegetation with mainly Festuca ovina s.l., Deschampsia flexuosa and reindeer lichens (Cladonia subgen. Cladina), proved to be rather stable. Since 1996 the lichen steppes occurring in biotope 2 were increasingly grass-encroached. Where some sand was still blowing in, lichen diversity did not change much, but gradually C. introflexus increased. However, without sand blowingin, a decreasing lichen cover and loss of some Red List lichens occurred. In general, in this grass-rich vegetation Calluna vulgaris will germinate and gradually a dry heath might develop. However, where dynamics had stopped and Pinus sylvestris seedlings established, succession to a young forest started. Vegetation change from open sand to lichen-rich vegetation is clearly related to a decrease in pH, an increase in organic matter, in % total N and in % total P. The lichen composition is clearly related to this gradient. The applied restoration measures on steep sand dunes in the eastern area, both of cutting pine trees and removing topsoil down to the mineral soil layer, proved to be successful. The sand kept drifting and the pioneer community thriving, including the lichen Stereocaulon condensatum characteristic of pioneer conditions, while the neophytic moss hardly increased between 1996 and 2006.
With increasing distribution of endovascular stroke therapies, transient middle cerebral artery occlusion (tMCAO) in mice now more than ever depicts a relevant patient population with recanalized M1 occlusion. In this case, the desired therapeutic effect of blood flow restauration is accompanied by breakdown of the blood-brain barrier (BBB) and secondary reperfusion injury. The aim of this study was to elucidate short and intermediate-term transcriptional patterns and the involved pathways covering the different cellular players at the neurovascular unit after transient large vessel occlusion. To achieve this, male C57Bl/6J mice were treated according to an intensive post-stroke care protocol after 60 min occlusion of the middle cerebral artery or sham surgery to allow a high survival rate. After 24 h or 7 days, RNA from microvessel fragments from the ipsilateral and the contralateral hemispheres was isolated and used for mRNA sequencing. Bioinformatic analyses allowed us to depict gene expression changes at two timepoints of neurovascular post-stroke injury and regeneration. We validated our dataset by quantitative real time PCR of BBB-associated targets with well-characterized post-stroke dynamics. Hence, this study provides a well-controlled transcriptome dataset of a translationally relevant mouse model 24 h and 7 days after stroke which might help to discover future therapeutic targets in cerebral ischemia/reperfusion injury.
This paper presents some of the major aspects of the history of German exile after 1933 and the history of the exile of German-speaking writers and intellectuals in Brazil. The second part of the title is focused on the works of exile written by Ulrich Becher and Hugo Simon in Brazil.
Der Beitrag befasst sich anhand von künstlerischen Arbeiten von Bettina Malcomess mit prekären Sichtbarkeiten und instabilen Narrativen im audiovisuellen Post-Apartheid-Archiv. Dabei werden das Nachwirken von Imperialismus und Apartheid in Südafrika und Strategien der Dekolonialisierung in künstlerischen Praktiken thematisiert.
In the title compound, C11H11N3O2, the dihedral angle between the central ethanone fragment and the 4-methoxyphenyl group is 2.9 (2)°, while that between the ethanone fragment and the triazole ring is 83.4 (2)°. The dihedral angle between the planes of the triazole and benzene rings is 81.7 (1)°. The 4-methoxyphenyl group is cis with respect to the ethanone fragment O atom across the exocyclic C—C bond. Das Ziel dieser Arbeit war die Evaluierung der Einsatzmöglichkeiten eines mikrostrukturierten Reaktorsystems in der heterogenen Katalyse. Hierzu wurde eine Reaktion herangezogen, welche typische Problemstellungen der heterogenen Katalyse abbildet. Zu diesen Problemen gehören Temperaturkontrolle, sichere Handhabung von explosiven Gasgemischen und das Erzielen von zufriedenstellenden Selektivitäten. Die Reaktion sollte außerdem bereits gut untersucht worden und die Prozessparameter aus der Literatur bekannt sein. Aus diesem Grund wurde die Partialoxidation von Ethen zu Ethenoxid an Silberkatalysatoren gewählt. Es konnte gezeigt werden, dass die Reaktion in einem Mikrostrukturreaktorsystem sicher durchführbar ist. Vor allem wurde an einer ganzen Reihe von Beispielen veranschaulicht, dass eine herausragende Eigenschaft des Mikrostrukturreaktors seine inhärente Explosionssicherheit ist. Gasgemische, welche sich mitten im explosiven Gemischbereich befanden, konnten bei Drücken von 2 bis 20 bar und Temperaturen von 230 bis 310 °C sicher gehandhabt werden. So konnte gezeigt werden, dass der Mikrostrukturreaktor sich dazu eignet Reaktionen mit explosiven Gasgemischen durchzuführen. Die Verwendung von Mikrostrukturreaktoren in der heterogenen Katalyse befindet sich noch im Anfangsstadium. Um Probleme bei der Übertragung von Katalysatorsystemen auf ein System mit Mikrostruktur zu vermeiden, erfolgte zunächst der Einsatz von Vollsilberkatalysatoren. Die Mikrostruktur wurde deshalb aus dem katalytisch aktiven Material selbst hergestellt. Die Herstellung wurde auf drei unterschiedliche Weisen (LIGA-, Ätz- und Sägeverfahren) durchgeführt. So konnte gezeigt werden, dass eine Kostenreduzierung bei der Darstellung von Mikrostrukturen möglich ist. Der Nachteil der Nutzung von Vollsilber war, dass sich deutlich schlechtere Selektivitäten bei der Partialoxidation von Ethen ergaben. Es konnte jedoch gezeigt werden, dass mit dem Mikrostrukturreaktor die Selektivitäten für Vollsilber im Schnitt 10 % über denen für Rohrreaktorexperimenten bei gleichen Umsätzen lagen. Die effektive Wärmeabführung und die homogene Verteilung der Wärme über den Mikrostrukturreaktor scheinen eine Verbesserung der Selektivität zu erbringen. Kinetische Untersuchungen zeigten, dass sowohl durch Anheben des Partialdrucks von Ethen als auch von Sauerstoff eine Erhöhung der Reaktionsgeschwindigkeit erzielt werden kann. Dabei wurde für Ethen eine formale Reaktionsordnung bei der Bildung von Ethenoxid von 0,53 gefunden, während sie für Sauerstoff 0,78 betrug. Mit diesen Untersuchungen wurde verdeutlicht, dass ein Erhöhen des Sauerstoffpartialdrucks einen positiven Einfluss auf die Selektivität hat. So konnte durch Anheben der Sauerstoffkonzentration von 5 %, wie es in industriellen Prozessen aus Sicherheitsgründen notwendig ist, auf bis zu 95 % eine Verbesserung der Selektivität von bis zu 15 % erzielt werden. Über diesen Sachverhalt wurde zwar bereits in der Literatur (16) berichtet, jedoch erfolgten die Untersuchungen hierfür unter Hochvakuumbedingungen. Der Mikrostrukturreaktor ermöglichte einen Nachweis dieses Phänomens auch unter Hochdruckbedingungen, wie sie für industrielle Reaktoren üblich sind. Damit konnte ein in der heterogenen Katalyse bekanntes Problem, nämlich die Übertragung von Erkenntnissen aus Ultrahochvakuumexperimenten auf Hochdruckbedingungen (pressure-gap), untersucht werden. Eine wissenschaftliche Prüfung, ob dem Ergebnis die gleichen Ursachen sowohl im Ultrahochvakuum als auch bei Hochdruckbedingungen zugrunde liegen, muss noch erfolgen. Es zeigte sich aber auch, dass durch eine Verweilzeiterhöhung keine weitere Verbesserung der Raum-Zeit-Ausbeute möglich ist. Vielmehr wurde klar, dass Reaktionsgeschwindigkeit und Selektivität mit längeren Verweilzeiten abnehmen. Als Grund hierfür konnte die Bildung von elementarem Kohlenstoff an der Silberoberfläche festgestellt werden. Aufgrund der Limitierung bei der Verweilzeit wurden maximale Umsätze von 24 % erzielt. Der Einsatz von 1,2-Dichlorethan als Oxidationsinhibitor für Vollsilber wurde ebenfalls untersucht. Dabei konnte die Selektivität auf bis zu 69 % gesteigert werden. Es erfolgte jedoch eine Einbuße an Aktivität von etwa 42 %. Es ist bekannt, dass die Oberflächenmorphologie von Silberkatalysatoren unter Reaktionsbedingungen starke Veränderungen erfährt. (68) Es wurde aufgezeigt, dass dies für die Oberfläche von mikrostrukturierten Silberfolien ebenfalls festzustellen ist. Dabei wurde gleichzeitig festgestellt, dass die Katalysatoren trotz unterschiedlicher Herstellungsmethoden und den daraus resultierenden unterschiedlichen Oberflächenmorphologien vergleichbare Aktivitäten aufweisen. Industriell verwendete Katalysatoren basieren auf alpha-Aluminiumoxid als Trägermaterial. Dabei wurde bereits seit vielen Jahren an Optimierungen des Katalysators gearbeitet. Durch das Einstellen der spezifischen Oberfläche und Partikelgröße des Silbers und den Einsatz von Alkali- und Erdalkalimetallen als Promotoren werden so Katalysatoren hergestellt, welche eine Selektivität von 80 % besitzen. Die Übertragung dieser Erkenntnisse auf ein Mikrostrukturreaktorsystem kann nicht ohne weiteres vorgenommen werden. Es wurden verschiedene Darstellungsmöglichkeiten für eine alpha-Aluminiumoxidschicht in einem Mikrostrukturreaktor untersucht. Dabei zeigte sich, dass nur die direkte Darstellung von alpha- Aluminiumoxid ohne Phasenumwandlung aus anderen Modifikationen erfolgversprechend ist. Eine Darstellung der Aluminiumoxidschicht durch Sol-Gel- oder CVD-Prozesse war nicht erfolgreich, da die für die Phasenumwandlung von gamma-Aluminiumoxid nach alpha-Aluminiumoxid notwendige Temperatur von 1100 °C die Ausbildung einer Eisenoxidschicht an der Oberfläche der mikrostrukturierten Edelstahlfolien zur Folge hatte. Diese eignete sich nicht als Träger. Alternativ wurde erfolgreich der Einsatz von aluminiumhaltigen Edelstählen untersucht. Diese bilden beim Ausheizen bei 1100 °C eine alpha-Aluminiumoxidschicht an der Oberfläche aus, welche mittels Sputtern mit Silber geträgert wurde. Katalytische Untersuchungen zeigten, dass mit dem Einsatz von alpha-Aluminiumoxidträgern eine Verbesserung der Selektivität im Vergleich zu Vollsilber von 17 % erreicht werden kann. Gleichzeitig konnte anhand eines Gegenüberstellens von katalytischen Daten mit TEM-Aufnahmen der Sputterschichten festgestellt werden, dass eine geschlossene Silberschicht an der Oberfläche notwendig ist, um eine zufriedenstellende Aktivität und Selektivität zu erzielen. Während bei Schichtdicken von 1 nm noch einzelne Silberinseln an der Oberfläche zu finden sind, liegt bei einer Schichtdicke von 5 nm eine fast geschlossene Silberschicht vor. Ein Anheben der Schichtdicke ergab keine weitere Verbesserung der Aktivität oder Selektivität. Dagegen ergab der Einsatz von 1,2-Dichlorethan eine weitere Steigerung der Selektivität auf 77 %. Industriell eingesetzte Rohrbündelreaktoren erreichen im Sauerstoffverfahren eine Selektivität von 80 %. Die hier erzielten 77 % Selektivität bei vergleichbaren Umsätzen zeigt, dass der Einsatz eines Mikrostrukturreaktors für die Synthese von Ethenoxid möglich ist, vor allem unter dem Gesichtspunkt, dass Potenzial für die Optimierung von Reaktoren und die Katalysatorpräparation besteht. Die Nutzung von Reaktionsbedingungen, wie Ethen in reinem Sauerstoff, und der daraus resultierenden Verbesserung für Aktivität und Selektivität, ermöglichen Raum-Zeit-Ausbeuten, die über denen von Industriereaktoren liegen. Ob Mikrostrukturreaktoren in industriellen Prozessen jemals eingesetzt werden, hängt allein von ökonomischen Faktoren ab. Dazu müsste die Selektivität über die bestehenden 80 % angehoben werden. Zur Zeit entfallen 80 % der Produktionskosten von Ethenoxid auf den Rohstoff Ethen, so dass jeder Prozentpunkt, um den die Selektivität angehoben werden könnte, eine deutliche Kosteneinsparung mit sich brächte und darüber entschiede, ob ein neuer Prozess eingeführt wird. Hierzu wäre es auch notwendig, die Kosten für die Produktion der Mikrostrukturreaktoren pro Volumeneinheit um mehrere Größenordnungen zu reduzieren. Außerdem müssten Lösungen entwickelt werden, welche die Peripherie des Reaktors betreffen, vor allem die Heizung und die Gasversorgung. Im Rahmen dieser Arbeit sollte überprüft werden, welche Leistungsfähigkeit ein Mikrostrukturreaktorprozess im Vergleich zu einem bestehenden Prozess besitzt. Es konnte dargestellt werden, dass Raum-Zeit-Ausbeuten über denen eines Industriereaktors erzielt werden können bei vergleichbareren Selektivitäten. Außerdem konnte gezeigt werden, dass der Mikrostrukturreaktor ein geeignetes Werkzeug ist, welches helfen kann, Reaktionen unter bisher nicht einfach zugänglichen Bedingungen durchzuführen.
Atovaquone is a substituted 2-hydroxynaphthoquinone that is used therapeutically to treat Plasmodium falciparum malaria, Pneumocystis carinii pneumonia, and Toxoplasma gondii toxoplasmosis. It is thought to act on these organisms by inhibiting the cytochrome bc1 complex. We have examined the interaction of atovaquone with the bc1 complex isolated from Saccharomyces cerevisiae, a surrogate, nonpathogenic fungus. Atovaquone inhibits the bc1 complex competitively with apparent Ki = 9 nm, raises the midpoint potential of the Rieske iron-sulfur protein from 285 to 385 mV, and shifts the g values in the EPR spectrum of the Rieske center. These results indicate that atovaquone binds to the ubiquinol oxidation pocket of the bc1 complex, where it interacts with the Rieske iron-sulfur protein. A computed energy-minimized structure for atovaquone liganded to the yeast bc1 complex suggests that a phenylalanine at position 275 of cytochrome b in the bovine bc1 complex, as opposed to leucine at the equivalent position in the yeast enzyme, is responsible for the decreased sensitivity of the bovine bc1 complex (Ki = 80 nm) to atovaquone. When a L275F mutation was introduced into the yeast cytochrome b, the sensitivity of the yeast enzyme to atovaquone decreased (Ki = 100 nm) with no loss in activity, confirming that the L275F exchange contributes to the differential sensitivity of these two species to atovaquone. These results provide the first molecular description of how atovaquone binds to the bc1 complex and explain the differential inhibition of the fungal versus mammalian enzymes.
A large reef manta ray (Manta alfredi) aggregation has been observed off the north Sudanese Red Sea coast since the 1950s. Sightings have been predominantly within the boundaries of a marine protected area (MPA), which was designated a UNESCO World Heritage Site in July 2016. Contrasting economic development trajectories have been proposed for the area (small-scale ecotourism and large-scale island development). To examine space-use, Wildlife Computers® SPOT 5 tags were secured to three manta rays. A two-state switching Bayesian state space model (BSSM), that allowed movement parameters to switch between resident and travelling, was fit to the recorded locations, and 50% and 95% kernel utilization distributions (KUD) home ranges calculated. A total of 682 BSSM locations were recorded between 30 October 2012 and 6 November 2013. Of these, 98.5% fell within the MPA boundaries; 99.5% for manta 1, 91.5% for manta 2, and 100% for manta 3. The BSSM identified that all three mantas were resident during 99% of transmissions, with 50% and 95% KUD home ranges falling mainly within the MPA boundaries. For all three mantas combined (88.4%), and all individuals (manta 1–92.4%, manta 2–64.9%, manta 3–91.9%), the majority of locations occurred within 15 km of the proposed large-scale island development. Results indicated that the MPA boundaries are spatially appropriate for manta rays in the region, however, a close association to the proposed large-scale development highlights the potential threat of disruption. Conversely, the focused nature of spatial use highlights the potential for reliable ecotourism opportunities.
Hintergrund: Einer der wichtigsten Gründe für das Therapieversagen von Zytostatika ist die MDR von malignen Zellen. Deren Modulation mit sogenannten Chemosen sitizern scheint ein erfolgversprechender Therapieansatz zu sein, so daß zahlreiche in vitro Studien initiiert wurden. Jedoch beachten diese Studien nur selten den Effekt der Proteinbindung auf die zelluläre Aufnahme von Zytostatika und der Wirkung von Chemosensitizern. Der Vorhersagewert der in vitro Tests für die in vivo Effektivität steht somit zur Diskussion. Die Ziele dieser Untersuchung waren es deshalb a) die Bindungscharakteristiken des Anthrazyklins IDA in HS und Zellkulturlösungen zu bestimmen, b) den Effekt der Proteinbindung, sowie des Chemosensitizers RVRP und des AkutPhaseProteins AGP, auf die Aufnahme und Retention von IDA in eine humane HL60 Leukämiezellinie zu untersuchen und c) die Bedeutung der Proteinbin dung für die in vivoAktivität von Zytostatika und Chemosensitizern zu analysieren. Methoden: Die Proteinbindung von IDA wurde mit Hilfe der Gleichgewichtsdialyse in HS und gebräuchlichen Zellkulturlösungen bestimmt. Die Zellaufnahme und Retention von IDA wurde in einer sensiblen und einer Pgpexprimierenden, resistenten humanen Leukämiezellinie (HL60 und HL60Vinc), sowie in Lymphozyten gesunder Probanden untersucht. Die Versuchsbedingungen wurden durch verschiedene Inkubationsmedien und die Zugabe von RVRP und AGP variiert. Die IDA Konzentration wurde spektrophotofluorometrisch analysiert. Ergebnisse: In den untersuchten Inkubationsmedien variierte die freie Fraktion von IDA um mehr als 7fach von ungefähr 60% in 15% FCS/PBS zu nur 8% in HS. In den Zellinien HL60 und HL60Vinc war die zelluläre Aufnahme von IDA in dem Medium mit niedrigen Proteinkonzentrationen ungefähr dreimal höher als in HS. Die Bedeutung von Pgp für das Resistenzverhalten der Leukämiezellinie zeigte sich durch die erniedrigte Akkumulation von IDA in den resistenten Zellen. RVRP konnte in allen Medien die Differenz der IDA Aufnahme und Retention zwischen der sensiblen und der resistenten Zellinie aufheben. Jedoch konnte RVRP nicht den Effekt von hohen Proteinkonzentrationen auf die IDA Aufnahme ausgleichen. In Anwesenheit von AGP konnte sowohl in den Medien mit hohen als auch niedrigen Protein konzentrationen nur eine sehr geringe Chemosensitizerwirkung nachgewiesen werden. Das Akkumulationsverhalten der Lymphozyten glich der sensiblen HL60 Zellinie. Schlußfolgerung: Proteinbindungsphänomene können die Aktivität von Zytostatika und Chemosensitizern erheblich beeinflussen. So ist in vitro die Serumproteinbindung eine wichtige Determinante für die zelluläre Aufnahme von IDA. Der Modulationseffekt von RVRP reagiert weniger sensitiv auf Unterschiede in der Gesamtproteinkonzentration, allerdings kann er durch erhöhte AGP Konzentrationen, wie sie bei Tumorpatienten vorkommen, erheblich beeinflußt werden. Bei der Interpretation von in vitro funktionellen Tests der zytostatischen Effektivität und Chemosensitizerwirkung sollte daher die Serumproteinbindung berücksichtigt und Untersuchungen in HS einbezogen werden.
Invasive fungal infections are associated with increased mortality in hematological patients. Despite considerable advances in antifungal therapy, the evaluation of suspected treatment failure is a common clinical challenge requiring extensive diagnostic testing to rule out potential causes, such as mixed infections. We present a 64-year-old patient with secondary AML, diabetes mellitus, febrile neutropenia, and sinusitis. While cultures from nasal tissue grew Aspergillus flavus, a microscopic examination of the tissue was suggestive of concomitant mucormycosis. However, fluorescence in situ hybridization (FISH) using specific probes targeting Aspergillus and Mucorales species ruled out mixed infection. This was confirmed by specific qPCR assays amplifying the DNA of Aspergillus, but not of Mucorales. These results provided a rational basis for step-down targeted therapy, i.e., the patient received posaconazole after seven days of calculated dual therapy with liposomal amphotericin B and posaconazole. Despite clinical response to the antifungal therapy, he died due to the progression of the underlying disease within two weeks after diagnosis of fungal infection. Molecular diagnostics applied to tissue blocks may reveal useful information on the etiology of invasive fungal infections, including challenging situations, such as with mixed infections. A thorough understanding of fungal etiology facilitates targeted therapy that may improve therapeutic success while limiting side effects.

Abstract
Objectives: Patients with open pulmonary tuberculosis (opTB) are subject to strict isolation rules. Sputum smear microscopy is used to determine infectivity, but sensitivity is lower than for culture. This study aimed to investigate the clinical relevance of this mismatch in contemporary settings.
Methods: Differential results between microscopy and culture were determined at the time of microscopic sputum conversion, from all patients with opTB between 01/2013 and 12/2017. In addition, data on HIV, multi/extensive drug-resistant TB status, time to smear- and cultural-negativity conversion were analyzed; and a Kaplan-Meier curve was developed.
Results: Of 118 patients with opTB, 58 had demographic data available for microbiological and clinical follow-up analysis; among these, 26 (44.8%) had still at least one positive culture result. Median time from opTB-treatment initiation to full microscopic sputum- or culture conversion, was 16.5 days (range 2-105), and 20 days (1-105), respectively (median difference: +3.5 days). Sixteen days after de-isolation, >90% had converted culturally. HIV- or multi/extensive drug-resistant TB status did not impact conversion time.
Conclusion: When patients with opTB were de-isolated after 3 negative sputum smear microscopy tests, a substantial part still revealed cultural growth of Mycobacterium tuberculosis complex, but it remains unclear, whether smear-negative and culturally-positive individuals on therapy are really infective. Thus, the clinical relevance of this finding warrants further investigation.
The taxonomic status of Alcyonium aurantiacum Quoy & Gaimard, 1833, an octocoral endemic to New Zealand, was reviewed through morpho-molecular data comparisons in an integrative approach. Molecular phylogenetic analyses (nuclear 28S and mitochondrial mtMutS) resolved New Zealand taxa as more closely related to other genera and nominal Alcyonium Linnaeus, 1758 from South America than to the genus’ North Atlantic type species. Due to low genetic variation, species delimitation relied predominantly on identifying consistent differences in sclerite and colony morphology. The former A. aurantiacum is reassigned to Kotatea gen. nov. as K. aurantiaca gen. et comb. nov. and seven new species are described in this genus (K. amicispongia gen. et sp. nov., K. lobata gen. et sp. nov., K. kapotaiora gen. et sp. nov., K. kurakootingotingo gen. et sp. nov., K. niwa gen. et sp. nov., K. raekura gen. et sp. nov., and K. teorowai gen. et sp. nov.). Three new species in Ushanaia gen. nov. are also described (U. ferruginea gen. et sp. nov., U. fervens gen. et sp. nov. and U. solida gen. et sp. nov. ). These descriptions increase our understanding of New Zealand’s endemic octocoral diversity and contribute to ongoing systematic revisions of Alcyonium.
Identifizierung und Charakterisierung von Liganden für Faktor VIII neutralisierende Antikörper

Das Fehlen von funktionellem Blutgerinnungsfaktor VIII (FVIII) in Hämophilie A- (HA-) Patienten wird durch Substitution mit FVIII-Präparaten therapiert. Die wesentlichste gegenwärtige Komplikation der FVIII-Ersatz-Therapie besteht in dem Auftreten von FVIII neutralisierenden Antikörpern (Inhibitoren) gegenüber exogenem FVIII. Diese können mittels verschiedener, kostenintensiver Therapien zur Induktion einer Immuntoleranz (ITI) mit unterschiedlichem Erfolg eliminiert werden. Für Patienten mit persistierenden Inhibitoren bedeuten diese nicht nur eine drastische Verminderung der Lebensqualität sondern ein lebensbedrohliches Szenario. Eine Liganden-vermittelte Blockierung von neutralisierenden anti-FVIII Antikörpern sowie die zielgerichtete Ansteuerung des Rezeptors FVIII-spezifischer Gedächtnis-B-Zellen stellen mögliche Ansätze zur Verwirklichung antigenspezifischer ITI-Strategien für eine dauerhafte, vollständige Eliminierung von FVIII-Inhibitoren dar. Zu diesem Zweck wurden in dieser Arbeit durch Screening von phagenpräsentierten, randomisierten Peptidbibliotheken mit Inhibitor-positiven Patientenplasmen Peptidliganden selektioniert. Diese wiesen eine spezifische Bindung von anti-FVIII Antikörpern in den verwendten Plasmen auf. Durch den Einsatz entsprechender Software konnten AS-Konsensusmotive der Peptidsequenzen möglichen, konformationellen, funktionellen Inhibitorepitopen in der A2- sowie C2-Domäne von FVIII zugeordnet werden. Die von in silico-Analysen vorgegebene Domänenspezifität der anti-FVIII Antikörper wurde in Bindungsstudien mit rekombinant exprimierten FVIII-Domänen verfiziert. Die korrespondierenden, synthetischen Peptidliganden blockierten die IgG-Bindung an FVIII und regenerierten partiell dessen Aktivität im Plasma. Die Peptide stellten funktionelle Mimotope der möglichen Inhibitorepitope in der A2- und C2-Domäne dar. Da FVIII neutralisierende Antikörper zumeist Epitope in beiden Domänen erkennen, wurden die Mimotope kombiniert, was in einer noch effektiveren Blockierung von FVIII-Inhibitoren resultierte. Weiterhin wiesen Mimotopkombinationen Kreuzreaktivität mit anti-FVIII IgG in heterologen Patientenplasmen auf. Durch Fusion der Peptide an die Multimerisierungsdomäne der alpha-Kette des humanen C4-Bindeproteins konnten in Zellkultur heptamere Proteine generiert werden. Gegenüber den synthetischen Peptiden wiesen die Multimere aufgrund ihrer Multivalenz sowie der strukturellen Integrität eine deutlich verbesserte Blockierung von anti-FVIII IgG auf. Das Multimerisierungskonzept erlaubte ferner die Kombination unterschiedlicher Peptidliganden in einem Heteromultimer, was anhand der selektierten, funktionellen Mimotope für mögliche A2- und C2-Epitope getestet wurde. Weiterhin zeichneten sich die Inhibitor-spezifischen Multimere gegenüber den synthetischen Peptiden durch deutlich verlängerte Halbwerstzeiten aus. In Präparationen peripherer mononuklearer Zellen (PBMCs) von Patienten färbten synthetische Peptide sowie Fluoreszenz-markierter FVIII B-Zellsubpopulationen mit einem Gedächntis-B-Zell Phänotyp (CD19+IgG+). Gedächtnis-B-Zellen in PBMCs wurden polyklonal stimuliert. Im ELISPOT-Verfahren konnten Tetanusspezifische, jedoch keine FVIII-spezifischen Zellen, detektiert werden. Im Gegensatz zu den verwandten Kontrollen bewirkte eine Präinkubation der Zellen mit dem Peptid 12C6, welches an das toxische D-AS-Peptid (KLAKLAK)2 gekoppelt war, allerdings eine Reduktion von anti-FVIII IgG in den Überständen stimulierter Zellen.

In this study, we investigate the composition of nucleated nanoparticles formed from these compounds in the CLOUD chamber experiments at CERN. The investigation is carried out via analysis of the particle hygroscopicity, ethanol affinity, oxidation state, and ion composition. Hygroscopicity was studied by a hygroscopic tandem differential mobility analyser and a cloud condensation nuclei counter, ethanol affinity by an organic differential mobility analyser and particle oxidation level by a high-resolution time-of-flight aerosol mass spectrometer. The ion composition was studied by an atmospheric pressure interface time-of-flight mass spectrometer. The volume fraction of the organics in the particles during their growth from sizes of a few nanometers to tens of nanometers was derived from measured hygroscopicity assuming the Zdanovski-Stokes-Robinson relationship, and compared to values gained from the spectrometers. The ZSR-relationship was also applied to obtain the measured ethanol affinities during the particle growth, which were used to derive the volume fractions of sulphuric acid and the other inorganics (e.g. ammonium salts). In the presence of sulphuric acid and ammonia, particles with a mobility diameter of 150 nm were chemically neutralised to ammonium sulphate. In the presence of oxidation products of pinanediol, the organic volume fraction of freshly nucleated particles increased from 0.4 to ∼0.9, with an increase in diameter from 2 to 63 nm. Conversely, the sulphuric acid volume fraction decreased from 0.6 to 0.1 when the particle diameter increased from 2 to 50 nm. The results provide information on the composition of nucleated aerosol particles during their growth in the presence of various combinations of sulphuric acid, ammonia, dimethylamine and organic oxidation products.

Diese Abschlussarbeit zur dialektalen Gliederung des Zazaki besteht aus 2 Hauptteilen. Teil 1 beschreibt nach kurzer Darlegung und Auswertung der bisherigen Forschung die dialektale Struktur der Sprache und die Einteilung in Hauptdialekte. Teil 2 besteht aus Übersetzungen eines Beispieltextes in verschiedene Dialekte und Mundarten sowie aus Sprachkarten mit Isoglossen.
Hypoxic pulmonary vasoconstriction (HPV) redistributes pulmonary blood flow from areas of low oxygen partial pressure to areas of normal or relativity high oxygen availability, thus optimising the matching of perfusion to ventilation and preventing arterial hypoxemia. Generalised alveolar hypoxia results in a sustained increase in pulmonary artery pressure which in turn leads to structural changes in the walls of the pulmonary vasculature (pulmonary vascular remodelling). Recent findings have indicated a role for cytochrome P450 (CYP) epoxygenase-derived epoxyeicosatrienoic acids (EETs) in hypoxia-induced pulmonary vasoconstriction. Given that the intracellular concentration of EETs is determined by the soluble epoxide hydrolase (sEH), which metabolises EETs to their less active dihydroxyeicosatrienoic acids (DHETs), we assessed the influence of the sEH and EETs on pulmonary artery pressure, acute and chronic HPV, and pulmonary vascular remodelling in the mouse lung. In isolated lungs from wild-type mice, acute HPV was significantly increased by sEH inhibition, an effect abolished by pre-treatment with CYP epoxygenase inhibitors and the EET antagonist 14,15-EEZE. The acute hypoxia-induced vasoconstriction and EET production were greater in lungs from sEH-/- mice than from wild-type mice and sEH inhibition had no further effect on HPV in lungs from the former animals, while MSPPOH (CYP epoxygenase inhibitor) and 14,15-EEZE decreased the response. Exogenous application of 11,12-EET increased pulmonary artery pressure in a concentration-dependent manner and enhanced acute HPV in wild-type lungs, while 14,15-EET and 11,12-DHET were without significant effect on pulmonary artery pressure. 5-HT2A receptor antagonism or Rho kinase inhibition shifted the EET concentration-response curve to the right and abrogated the EET- and sEH inhibition-induced potentiation of acute hypoxic vasoconstriction. In lungs from wild-type and sEH-/- mice, hypoxic preconditioning (hypoxic ventilation for 10 minutes) enhanced the 5-HT response. 1-Adamantyl-3-cyclohexylurea (ACU), a sEH inhibitor, further amplified the hypoxia-induced 5-HT-hypersensitivity in wild-type mice. However, after hypoxic preconditioning, the sEH-/- lungs displayed a striking leftward shift in the 5-HT response. 11,12-EET can activate TRPC6 channels in endothelial cells by eliciting its translocation to the plasma membrane, more specifically to membrane domains enriched with the caveolae marker caveolin-1. This effect was also observed in rat pulmonary artery smooth muscle cells overexpressing the channel. Exposure of the latter cells to acute hypoxia also stimulated the intracellular translocation of TRPC6 to caveolae, an effect that was sensitive to the EET antagonist. The EET-induced translocation of TRPC6 channels was prevented by a 5-HT2A receptor antagonist but not by a Rho kinase inhibitor. Moreover, while acute hypoxia and 11,12-EET increased pulmonary pressure in lungs from TRPC6+/- mice, lungs from TRPC6-/- mice did not respond to either stimuli. These results indicate that the sEH and CYP-derived EETs are involved in acute HPV and that EET-induced pulmonary contraction under normoxic and hypoxic conditions involves a TRPC6 channel, a 5-HT2A receptor-dependent pathway and Rho kinase activation. In the second part of the study the role of the sEH in the development of pulmonary hypertension and vascular remodelling induced in mice by exposure to hypoxia (10% O2) for 21 days was analysed. In wild-type mice, chronic hypoxia decreased the pulmonary expression/activity of the sEH, induced right heart hypertrophy and erythropoiesis, and increased the number of partially and fully muscularised pulmonary resistance arteries (by 3-fold). Moreover, in HEK 293 cells, hypoxia (1% O2 up to 24 h) decreased sEH promoter activity by 50%. In isolated lungs, pre-exposure to chronic hypoxia significantly increased baseline perfusion pressures and potentiated the acute HPV. While an sEH inhibitor, ACU, potentiated acute HPV in lungs from mice maintained in normoxic conditions, it had no effect on HPV in lungs from mice exposed to hypoxia. The EET antagonist, 14,15-EEZE, abolished the sEH inhibitor-dependent increase in acute HPV in normoxic lungs and decreased HPV in chronic hypoxic lungs. Hypoxia-induced right heart hypertrophy and erythropoiesis were more pronounced in sEH-/- than in wild-type mice. Under normoxic and hypoxic conditions the muscularisation of resistance pulmonary arteries was greater in lungs from sEH-/- mice than in lungs from wild-type mice. sEH-/- mice also displayed an enhanced acute HPV, compared to that observed in wild-type mice and chronic exposure to hypoxia did not further potentiate acute HPV. However, in the presence of 14,15-EEZE responses returned to levels observed in normoxic lungs from wild-type animals. Furthermore, immunohistochemistry demonstrated an extensive expression of the sEH in the medial wall of pulmonary arteries from human donor lungs. Whereas sEH expression was not detectable in samples from pulmonary hypertension patients, indicating that the sEH is involved in hypoxia-induced pulmonary vascular remodelling and hypoxic pulmonary vasoconstriction. Taken together, the results presented in this thesis indicate that the expression/activity of the sEH is an important determinant of the magnitude of acute and chronic hypoxia-induced pulmonary vasoconstriction and pulmonary vascular remodelling by inactivating vasoconstrictor CYP-derived EETs. As sEH inhibitors are currently being developed for the treatment of human systemic hypertension, it should be noted that these compounds may even promote the development of pulmonary hypertension.
We present results for calculating fusion cross-sections using a new microscopic approach based on a time-dependent density-constrained DFT calculations. The theory is implemented by using densities and other information obtained from TDDFT time-evolution of the nuclear system as a constraint on the density for DFT calculations.
İsviçreli gazeteci, foto muhabiri, yazar, fotoğrafçı Annemarie Schwarzenbach, gazetecilik göreviyle Ekim 1933'te, Irak'ta sonlanacak doğu yolculuğuna çıkmıştır. Seyahatinin iki ayını Türkiye'de geçirmiştir. Gördüklerini, edindiği izlenimler çerçevesinde kaleme alarak, 1935 yılında "Winter in Vorderasien - Tagebuch einer Reise" başlığıyla yayınlatmıştır. Seyahat, Türkiye'de reform çalışmalarına ve Cumhuriyet'in onuncu yılına denk gelmektedir. Ancak aktarımlarına bakıldığında, eleştirel bakış açısı dikkat çekmektedir. Yürütülecek bu çalışma ile Schwarzenbach'ın Alman edebiyatına sunduğu Türkiye imgesinin anlaşılmasına katkı sağlanması amaçlanmıştır. Bu nedenle, seyahatname Türkiye'ye dair tasvirleri çerçevesinde ele alınacaktır.
Fırtına ve Coşku edebi akımı yazarlarından Heinrich Leopold Wagner'in "Kindermörderin" adlı eseri, akımın en önemli eserleri arasında gösterilmektedir. Wagner, eseri yayınlandığında, yazım stili ve konuları açıklıkla ele almış olması nedeniyle oldukça ağır eleştirilere maruz kalmıştır. Burjuva trajedisi türündeki eserinde, 18. yüzyıl toplum yaşamının en önemli konularından olan çocuk cinayeti motifini işlemiştir. Ancak, motifin karakteristik özelliği olan soylu bir erkekle saf burjuva kızı ilişkisini kurgularken, motifin ayrıntılarında değişikliğe gittiği görülmektedir. Bu bağlamda, kısaca Fırtına ve Coşku akımına ve 18. yüzyıl Alman toplumunda çocuk cinayeti olgusuna değinilecektir. Eser, dönem ve olguların arka planına dair verilen bu bilgilerden sonra, Fırtına ve Coşku akımının çocuk cinayeti motifini işleyişi ele alınacaktır. Ardından, Wagner'in, eserinde çocuk cinayeti motifini işlerken gittiği değişiklikler ve bu yolla ortaya koyduğu olgular saptanacaktır. Böylece, yürütülecek bu çalışmayla, çocuk cinayeti motifi konusunda yapılmış genel geçer tanımların dışına çıktığı ortaya konularak, Wagner'in eserine ve çocuk cinayeti motifi üzerine yapılacak araştırmalara katkı sağlanması amaçlanmıştır.
Epoxides and diols of polyunsaturated fatty acids (PUFAs) are bioactive and can influence processes such as tumor cell proliferation and angiogenesis. Studies with inhibitors of the soluble epoxide hydrolase (sEH) in animals overexpressing cytochrome P450 enzymes or following the systemic administration of specific epoxides revealed a markedly increased incidence of tumor metastases. To determine whether PUFA epoxides increased metastases in a model of spontaneous breast cancer, sEH-/- mice were crossed onto the polyoma middle T oncogene (PyMT) background. We found that the deletion of the sEH accelerated the growth of primary tumors and increased both the tumor macrophage count and angiogenesis. There were small differences in the epoxide/diol content of tumors, particularly in epoxyoctadecamonoenic acid versus dihydroxyoctadecenoic acid, and marked changes in the expression of proteins linked with cell proliferation and metabolism. However, there was no consequence of sEH inhibition on the formation of metastases in the lymph node or lung. Taken together, our results confirm previous reports of increased tumor growth in animals lacking sEH but fail to substantiate reports of enhanced lymph node or pulmonary metastases.
Der Name Engelbert Humperdinck wird von den meisten Musikinteressierten unwillkürlich mit seinem berühmtesten Werk "Hänsel und Gretel" oder allgemein mit dem Begriff "Märchenoper" assoziiert. Auch herausragende Musikwissenschaftler - wie z.B. Carl Dahlhaus im "Neuen Handbuch der Musikwissenschaft" - beschränken sich beim Stichwort "Humperdinck" in der Regel auf Erläuterungen zu "Hänsel und Gretel". Humperdinck hat in der Tat eine ganze Reihe von Märchenstoffen vertont: Er begann mit Liedern zu Märchenspielen, die seine Schwester Adelheid Wette für ihre Kinder geschrieben hatte ("Schneewittchen" 1888, "Hänsel und Gretel" 1890 und "Die sieben Geislein" 1895). Nach dem Erfolg der durchkomponierten Oper "Hänsel und Gretel" 1893 versuchte er sich an der Gattung Melodram ("Königskinder" 1895/97 nach einem Text von Elsa Bernstein), einer Mischform aus Singspiel und Melodram ("Dornröschen" 1902 nach einem Text von Elisabeth Filhés-Ebeling) und schließlich wieder an der durchkomponierten Märchenoper ("Königskinder" 1910). Außerdem schrieb er ein Vorspiel und zwei Musiknummern zu Maurice Maeterlincks "Der blaue Vogel" (1910-12).
Jedoch war dies nur ein Bereich der Musik für die Bühne, in dem er sich betätigte. Eine bedeutende Phase war für Humperdinck die Zusammenarbeit mit Max Reinhardt am Deutschen Theater und Alfred Halm am "Deutschen Schauspielhaus" in Berlin (von 1905 bis 1907), in der u.a. Bühnenmusik zu vier Shakespeareschen Werken entstand ("Der Kaufmann von Venedig", "Der Sturm", "Das Wintermärchen" und "Was ihr wollt"). In seiner letzten Schaffensperiode konnte Humperdinck sich verstärkt der Gattung widmen, die ihm schon vor Hänsel und Gretel als Ausweg aus der Wagner-Nachfolge-Lethargie erschienen war, der komischen Oper ("Heirat wider Willen" 1905, "Die Marketenderin" 1914, "Gaudeamus" 1919).
In diesem Beitrag soll "Dornröschen" genauer betrachtet werden - ein Märchenspiel, das im Unterschied zu den beiden Märchenopern Humperdincks wenig Erfolg hatte und in der Literatur bisher so gut wie gar nicht behandelt wurde. Wie ist das Werk entstanden, was ist das Besondere an ihm, und welche Gründe gibt es für sein Scheitern auf der Bühne? Zunächst soll je doch ein kurzer Überblick über Inhalt, Besetzung und Form in das Werk ein führen...
„Zu viel Musik“ lautet der Titel eines Aufsatzes des Pianisten, Komponisten, Dirigenten und Musikschriftstellers Ferdinand Hiller (1811-1885) über die Musikbelästigung eines musikalischen Bürgers durch unterschiedlichste Quellen in seinem Umfeld im Laufe eines Tages, sicherlich auf eigenen Erfahrungen fußend.1 Er wird als Titel für diesen Beitrag im übertragenen Sinne zitiert, weil der Nachlass Hiller in der Universitätsbibliothek Frankfurt am Main beim ersten Sichten bisher sehr unübersichtlich erschien. Der Nachlass umfasst 74 Bände Musikhandschriften, davon 21 Sammelhandschriften, dazu 9 Mappen mit Stimmen, 59 Bände Musikdrucke eigener Werke, davon 29 Sammelbände, dazu eine dicke Mappe mit Stimmen. Weitere Handschriften wurden nachträglich erworben. In verschiedenen Sammlungen sind darüber hinaus Briefe und Bildnisse Hillers überliefert.
Dieser Beitrag hat das Ziel, eine strukturierte Übersicht über die enthaltenen Werke zu geben: Der Inhalt der im Druck überlieferten Sammelbände wird anhand einer Tabelle dargestellt. Den Hauptteil dieses Beitrages bildet eine nach musikalischen Gattungen gegliederte Liste der in Frankfurt überlieferten Musikautographe, die als Register zu dem mittler weile digital verfügbaren Nachlassverzeichnis von Christine Ihl genutzt werden kann.
Zwei große Theaterzettelsammlungen mit über 100.000 Stück sind seit dem ersten Drittel des 20. Jahrhunderts in der damaligen Stadt- und heutigen Universitätsbibliothek Frankfurt am Main beheimatet und machen sie in dieser Hinsicht zu einer bedeutenden theaterhistorischen Institution im deutschsprachigen Raum: die Sammlung der Städtischen Bühnen Frankfurt am Main und ihrer Vorläufer sowie die Sammlung des Manskopfschen Musik- und Theaterhistorischen Museums...
Exzellenz ist in der Frankfurter Universität nicht nur in den Bereichen Naturwissenschaften, Gesellschaftswissenschaften und Wirtschaftwissenschaften zu finden. Eine Reihe historischer Sammlungen gibt ihr eine einzigartige Prägung und hebt sie von anderen Universitäten ab. Der Zufall – ein Helfer und kein Störenfried : warum die Wissenschaft stochastische Modelle braucht

Der Zufall hat in den Wissenschaften weithin einen zweifelhaften Ruf. Für die Philosophie hat Hegel festgestellt: »Die philosophische Betrachtung hat keine andere Absicht, als das Zufällige zu entfernen« (Die Vernunft in der Geschichte, 1822) – und ähnlich denkt man auch in anderen Wissenschaften. Die Auseinandersetzungen der Physik mit dem Zufall sind verschlungen und bis heute von Kontroversen begleitet. Was die Biologie betrifft, so herrscht noch einiger Argwohn gegenüber den modernen Evolutionstheorien, die sich entscheidend auf den Zufall stützen. Und dass derartige Theorien unvereinbar sind mit der Vorstellung von einer göttlichen Schöpfung der Welt, gilt unter manchen ihrer Gegner wie Befürworter als ausgemacht.
In this article, we build on research arguing that linguistic self-representation on social media can be viewed as a form of face-work and that the strategies employed by users are influenced by both a desire to connect with others and a need to preserve privacy. Drawing on our own analyses of usernames as well as that of others which were conducted as part of a large-scale project investigating usernames in 14 languages (Schlobinski/T. Siever 2018a), we argue that these conflicting goals of wanting to be recognised as an authentic member of an in-group while retaining a degree of anonymity are also observable in the choice of username. Online self-naming can thus be viewed as a key practice in the debate of face-work on social media platforms, because names and naming strategies can be studied more readily than broader and more complex aspects, such as stylistic variation or text-image interdependence, while at the same time forming part of these.
Exposure to community violence through witnessing or being directly victimized has been associated with conduct problems in a range of studies. However, the relationship between community violence exposure (CVE) and conduct problems has never been studied separately in healthy individuals and individuals with conduct disorder (CD). Therefore, it is not clear whether the association between CVE and conduct problems is due to confounding factors, because those with high conduct problems also tend to live in more violent neighborhoods, i.e., an ecological fallacy. Hence, the aim of the present study was: (1) to investigate whether the association between recent CVE and current conduct problems holds true for healthy controls as well as adolescents with a diagnosis of CD; (2) to examine whether the association is stable in both groups when including effects of aggression subtypes (proactive/reactive aggression), age, gender, site and socioeconomic status (SES); and (3) to test whether proactive or reactive aggression mediate the link between CVE and conduct problems. Data from 1178 children and adolescents (62% female; 44% CD) aged between 9 years and 18 years from seven European countries were analyzed. Conduct problems were assessed using the Kiddie-Schedule of Affective Disorders and Schizophrenia diagnostic interview. Information about CVE and aggression subtypes was obtained using self-report questionnaires (Social and Health Assessment and Reactive-Proactive aggression Questionnaire (RPQ), respectively). The association between witnessing community violence and conduct problems was significant in both groups (adolescents with CD and healthy controls). The association was also stable after examining the mediating effects of aggression subtypes while including moderating effects of age, gender and SES and controlling for effects of site in both groups. There were no clear differences between the groups in the strength of the association between witnessing violence and conduct problems. However, we found evidence for a ceiling effect, i.e., individuals with very high levels of conduct problems could not show a further increase if exposed to CVE and vice versa. Results indicate that there was no evidence for an ecological fallacy being the primary cause of the association, i.e., CVE must be considered a valid risk factor in the etiology of CD.
Background: In Europe, the number of females exhibiting oppositional defiant disorder (ODD) and conduct disorder (CD) is growing. Many of these females live in youth welfare institutions. Consequently, there is a great need for evidence-based interventions within youth welfare settings. A recently developed approach targeting the specific needs of girls with ODD and CD in residential care is START NOW. The aim of this group-based behavioural skills training programme is to specifically enhance emotional regulation capacities to enable females with CD or ODD to appropriately deal with daily-life demands. It is intended to enhance psychosocial adjustment and well-being as well as reduce oppositional and aggressive behaviour. We present the study protocol (version 4.1; 10 February 2016) of the FemNAT-CD intervention trial titled "Group-Based Treatment of Adolescent Female Conduct Disorders: The Central Role of Emotion Regulation".
Methods/design: The study is a prospective, confirmatory, cluster-randomised, parallel-group, multi-centre, randomised controlled trial with 128 institutionalised female adolescents who fulfil the diagnostic criteria of ODD and/or CD. Institutions/wards will be randomised either to provide the 12-week skills training as an add-on intervention or to provide treatment as usual. Once the first cycle is completed, each institution will run a second cycle with the opposite condition. Primary endpoints are the pre-post change in number of CD/ODD symptoms as assessed by a standardised, semi-structured psychiatric interview (Kiddie Schedule for Affective Disorders and Schizophrenia for School-Age Children–Present and Lifetime, CD/ODD section) between baseline and the end of intervention, as well as between baseline and a 3-month follow-up point. Secondary objectives include pre-post change in CD/ODD-related outcome measures, most notably emotional regulation on a behavioural and neurobiological level after completion of START NOW compared with treatment as usual.
Discussion: To our knowledge, this study is the first to date to systematically investigate the effectiveness of an adapted integrative psychosocial intervention designed for female adolescents with ODD and CD in youth welfare settings.

Kurzfassung eines Vortrags gehalten auf der GMA-Tagung vom 04.11.-06.11.2005 in Münster. Aufgrund wenig überzeugender Leistungen seiner Studierenden im vorklinischen Studienabschnitt hat der Fachbereich Medizin der Goethe-Universität zum Wintersemester 2001/02 eine völlig neue Studienordnung eingeführt, deren wesentliche Ziele das erfolgreiche Erlernen natur-, geistes- und sozialwissenschaftlicher Komponenten der normalen Biologie des Menschen mit optimaler Vorbereitung auf den klinischen Studienabschnitt binnen zweier Studienjahre und die Kompatibilität mit der ÄAppO und der ZÄAppO (jeweils alte und neue Version bzw. Vorentwurf) sind. Grundprinzipien des Curriculums sind: 1. Logisch aufeinander abgestimmte Themenblöcke, 2. Integration von makroskopischer und mikroskopischer Anatomie, 3. Interdisziplinärität der Themenblöcke, 4. Immanente Wiederholung von Lehrinhalten durch enge Verzahnung von Plenar- und Kleingruppenveranstaltungen, 5. Aufteilung der Studienanfänger in gemischte Gruppen nach den Kriterien Geschlecht, Herkunft und Studienfach, 6. Erlernen des Umgangs mit Patienten schon vom ersten Semester an durch "Anatomie am Lebenden"-Kurse und durch den Kurs Einführung in die klinische Medizin. Wichtigste Ergebnisse: Steigerung der Referenzgruppe beim Physikum von ca. 25% auf >60%, relativ guter Prüfungserfolg, hervorragende Sozialisation durch die frühe randomisierte Zuordnung in Kleingruppen und der sehr glatte Übergang in den klinischen Abschnitt des Medizin- bzw. Zahnmedizinstudiums.
Introduction: Preclinical CT-guided radiotherapy platforms are increasingly used but the CT images are characterized by poor soft tissue contrast. The aim of this study was to develop a robust and accurate method of MRI-guided radiotherapy (MR-IGRT) delivery to abdominal targets in the mouse.
Methods: A multimodality cradle was developed for providing subject immobilisation and its performance was evaluated. Whilst CT was still used for dose calculations, target identification was based on MRI. Each step of the radiotherapy planning procedure was validated initially in vitro using BANG gel dosimeters. Subsequently, MR-IGRT of normal adrenal glands with a size-matched collimated beam was performed. Additionally, the SK-N-SH neuroblastoma xenograft model and the transgenic KPC model of pancreatic ductal adenocarcinoma were used to demonstrate the applicability of our methods for the accurate delivery of radiation to CT-invisible abdominal tumours.
Results: The BANG gel phantoms demonstrated a targeting efficiency error of 0.56 ± 0.18 mm. The in vivo stability tests of body motion during MR-IGRT and the associated cradle transfer showed that the residual body movements are within this MR-IGRT targeting error. Accurate MR-IGRT of the normal adrenal glands with a size-matched collimated beam was confirmed by γH2AX staining. Regression in tumour volume was observed almost immediately post MR-IGRT in the neuroblastoma model, further demonstrating accuracy of x-ray delivery. Finally, MR-IGRT in the KPC model facilitated precise contouring and comparison of different treatment plans and radiotherapy dose distributions not only to the intra-abdominal tumour but also to the organs at risk.
Conclusion: This is, to our knowledge, the first study to demonstrate preclinical MR-IGRT in intra-abdominal organs. The proposed MR-IGRT method presents a state-of-the-art solution to enabling robust, accurate and efficient targeting of extracranial organs in the mouse and can operate with a sufficiently high throughput to allow fractionated treatments to be given.
We here report the complete nucleotide sequence of the 47.9 kb mitochondrial (mt) genome from the obligate aerobic yeast Yarrowia lipolytica. It encodes, all on the same strand, seven subunits of NADH: ubiquinone oxidoreductase (ND1-6, ND4L), apocytochrome b (COB), three subunits of cytochrome oxidase (COX1, 2, 3), three subunits of ATP synthetase (ATP6, 8 and 9), small and large ribosomal RNAs and an incomplete set of tRNAs. The Y. lipolytica mt genome is very similar to the Hansenula wingei mt genome, as judged from blocks of conserved gene order and from sequence homology. The extra DNA in the Y. lipolytica mt genome consists of 17 group 1 introns and stretches of A+Trich sequence, interspersed with potentially transposable GC clusters. The usual mould mt genetic code is used. Interestingly, there is no tRNA able to read CGN (arginine) codons. CGN codons could not be found in exonic open reading frames, whereas they do occur in intronic open reading frames. However, several of the intronic open reading frames have accumulated mutations and must be regarded as pseudogenes. We propose that this may have been triggered by the presence of untranslatable CGN codons. This sequence is available under EMBL Accession No. AJ307410.
Interessiert man sich im Hinblick auf den Primitivismus für das Wechselverhältnis von Ethnologie, Kunstgeschichte und den Künsten, versprechen Carl Einsteins kunstwissenschaftliche Schrift 'Negerplastik' und sein kubistisch inspirierter Text 'Bebuquin oder die Dilettanten des Wunders' aufschlussreich für eine Untersuchung zu sein. 'Negerplastik' erörtert kunstgeschichtliche Fragestellungen zur Raumdarstellung am Verhältnis von kubistischer und primitiver Kunst und diskutiert letztere unter einem mit der Ethnologie geteilten Interesse an Kult und Ritual. 'Bebuquin oder die Dilettanten des Wunders' wiederum macht kubistische Formprinzipien und die zirzensischen Künste als Importe des Primitiven in die moderne europäische Großstadt lesbar und betreibt dadurch eine literarische Variante innerkultureller Ethnographie. Die Hypothese teilend, Intermedialität stelle für den modernen Primitivismus ein konstitutives Moment dar, möchte ich ein Medium des Primitivismus in den Fokus rücken, das kulturelle Transfers bei der Konzeptualisierung und Repräsentation des Primitiven besonders greifbar und transparent macht: die Puppe.

Die Rheumatoide Arthritis ist eine Autoimmunerkrankung, die vor allem die Gelenke befällt. Hierbei kommt es zu einer ausgeprägten Synovialitis unter Beteiligung von Tund B-Lymphozyten, NK-Zellen, Makrophagen, dendritischen Zellen sowie Synoviozyten vom Fibroblastentyp. Dies führt zur Zerstörung des Gelenkknorpels und schliesslich des Gelenkes selbst. Eine entscheidende Rolle bei der Ausbildung und Aufrechterhaltung der Entzündung spielen synoviale T-Zellen, die unter anderem eine gesteigerte Apoptoseresistenz zeigen. Der Insulin-like Growth Factor Type 1 Rezeptor wird auf aktivierten T-Zellen vermehrt exprimiert und führt zu einer vermehrten Proliferation und verminderten Apoptoseneigung. Insulin-like Growth Factor 2 ist ein wichtiger Ligand dieses Rezeptors und wird normalerweise nur vom väterlichen Allel transkribiert. Unter Loss of Imprinting von Insulin-like Growth Factor 2 versteht man die biallelische Expression dieses Wachstumsfaktors. LOI von IGF-2 wurde bei aktivierten, proliferienden T-Zellen beobachtet. In dieser Arbeit wurden Synovial Tissue Mononuclear Cells (STMC) aus Synovialisbiopsaten von Patienten mit aktiver RA auf ein LOI von IGF-2 hin untersucht, um herauszufinden ob LOI von IGF-2 eine Rolle bei der Entstehung und Aufrechterhaltung des lokalen Entzündungsgeschehens spielen könnte. Zusätzlich erfolgten Versuche mit Peripheral Blood Mononuclear Cells (PBMC) von gesunden Versuchspersonen, um mehr über die Zusammenhänge zwischen Stimulation, Proliferation und LOI von IGF-2 herauszufinden. Die Synovialisbiopsate stammten aus der orthopädischen Universitätsklinik, wo sie im Rahmen von gelenkchirurgischen Eingriffen bei RA-Patienten mit aktiver Synovialitis entnommen wurden. Alle untersuchten Biopsate zeigten ein Retention of Imprinting, also eine monoallelische Expression von IGF-2. Die Versuche mit PBMC konnten keinen direkten Zusammenhang zwischen Stimulation, Proliferation und LOI von IGF-2 zeigen, diese Ergebnisse sind mit neueren Untersuchungen vereinbar. Zusammenfassend lässt sich sagen, dass LOI von IGF-2 als Faktor für das Auftreten oder die Aufrechterhaltung einer RA keine Rolle zu spielen scheint.
