Background: The human chromosomal region 9p21.3 has been shown to be strongly associated with Coronary Heart Disease (CHD) in several Genome-wide Association Studies (GWAS). Recently, this region has also been shown to be associated with Aggressive Periodontitis (AgP), strengthening the hypothesis that the established epidemiological association between periodontitis and CHD is caused by a shared genetic background, in addition to common environmental and behavioural risk factors. However, the size of the analyzed cohorts in this primary analysis was small compared to other association studies on complex diseases. Using our own AgP cohort, we attempted to confirm the described associations for the chromosomal region 9p21.3. Methods: We analyzed our cohort consisting of patients suffering from the most severe form of AgP, generalized AgP (gAgP) (n = 130) and appropriate periodontally healthy control individuals (n = 339) by genotyping four tagging SNPs (rs2891168, rs1333042, rs1333048 and rs496892), located in the chromosomal region 9p21.3, that have been associated with AgP. Results: The results confirmed significant associations between three of the four SNPs and gAgP. The combination of our results with those from the study which described this association for the first time in a meta-analysis of the four tagging SNPs produced clearly lower p-values compared with the results of each individual study. According to these results, the most plausible genetic model for the association of all four tested SNPs with gAgP seems to be the multiplicative one. Conclusion: We positively replicated the finding of an association between the chromosomal region 9p21.3 and gAgP. This result strengthens support for the hypothesis that shared susceptibility genes within this chromosomal locus might be involved in the pathogenesis of both CHD and gAgP.
On demand treatment and home therapy of hereditary angioedema in Germany - the Frankfurt experience

Background: Manifestation of acute edema in hereditary angioedema (HAE) is characterized by interindividual and intraindividual variability in symptom expression over time. Flexible therapy options are needed. Methods: We describe and report on the outcomes of the highly individualized approach to HAE therapy practiced at our HAE center in Frankfurt (Germany). Results: The HAE center at the Frankfurt University Hospital currently treats 450 adults with HAE or AAE and 107 pediatric HAE patients with highly individualized therapeutic approaches. 73.9% of the adult patients treat HAE attacks by on-demand therapy with pasteurized pd C1-INH concentrate, 9.8% use additional prophylaxis with attenuated androgens, 1% of the total patient population in Frankfurt has been treated with Icatibant up to now. In addition adult and selected pediatric patients with a high frequency of severe attacks are instructed to apply individual replacement therapy (IRT) with pasteurized pd C1-INH concentrate. Improvement on Quality of Life items was shown for these patients compared to previous long-term danazol prophylaxis. Home treatment of HAE patients was developed in the Frankfurt HAE center in line with experiences in hemophilia therapy and has so far been implemented over a period of 28 years. At present 248 (55%) of the adult patients and 26 (24%) of the pediatric patients are practicing home treatment either as on demand or IRT treatment. Conclusions: In conclusion, the individualized home therapies provided by our HAE center, aim to limit the disruption to normal daily activities that occurs for many HAE patients. Furthermore, we seek to optimize the economic burden of the disease while offering a maximum quality of life to our patients.
Background We published the Canadian 2003 International Consensus Algorithm for the Diagnosis, Therapy, and Management of Hereditary Angioedema (HAE; C1 inhibitor [C1-INH] deficiency) and updated this as Hereditary angioedema: a current state-of-the-art review: Canadian Hungarian 2007 International Consensus Algorithm for the Diagnosis, Therapy, and Management of Hereditary Angioedema. Objective To update the International Consensus Algorithm for the Diagnosis, Therapy and Management of Hereditary Angioedema (circa 2010). Methods The Canadian Hereditary Angioedema Network (CHAEN)/Reseau Canadien d'angioedeme hereditaire (RCAH) (www.haecanada.com) and cosponsors University of Calgary and the Canadian Society of Allergy and Clinical Immunology (with an unrestricted educational grant from CSL Behring) held our third Conference May 15th to 16th, 2010 in Toronto Canada to update our consensus approach. The Consensus document was reviewed at the meeting and then circulated for review. Results This manuscript is the 2010 International Consensus Algorithm for the Diagnosis, Therapy and Management of Hereditary Angioedema that resulted from that conference. Conclusions Consensus approach is only an interim guide to a complex disorder such as HAE and should be replaced as soon as possible with large phase III and IV clinical trials, meta analyses, and using data base registry validation of approaches including quality of life and cost benefit analyses, followed by large head-to-head clinical trials and then evidence-based guidelines and standards for HAE disease management.
Background: The potential anti-cancer effects of mammalian target of rapamycin (mTOR) inhibitors are being intensively studied. To date, however, few randomised clinical trials (RCT) have been performed to demonstrate anti-neoplastic effects in the pure oncology setting, and at present, no oncology endpoint-directed RCT has been reported in the high-malignancy risk population of immunosuppressed transplant recipients. Interestingly, since mTOR inhibitors have both immunosuppressive and anti-cancer effects, they have the potential to simultaneously protect against immunologic graft loss and tumour development. Therefore, we designed a prospective RCT to determine if the mTOR inhibitor sirolimus can improve hepatocellular carcinoma (HCC)-free patient survival in liver transplant (LT) recipients with a pre-transplant diagnosis of HCC. Methods: The study is an open-labelled, randomised, RCT comparing sirolimus-containing versus mTOR-inhibitor-free immunosuppression in patients undergoing LT for HCC. Patients with a histologically confirmed HCC diagnosis are randomised into 2 groups within 4-6 weeks after LT; one arm is maintained on a centre-specific mTOR-inhibitor-free immunosuppressive protocol and the second arm is maintained on a centre-specific mTOR-inhibitor-free immunosuppressive protocol for the first 4-6 weeks, at which time sirolimus is initiated. A 3-year recruitment phase is planned with a 5-year follow-up, testing HCC-free survival as the primary endpoint. Our hypothesis is that sirolimus use in the second arm of the study will improve HCC-free survival. The study is a non-commercial investigator-initiated trial (IIT) sponsored by the University Hospital Regensburg and is endorsed by the European Liver and Intestine Transplant Association; 13 countries within Europe, Canada and Australia are participating. Discussion: If our hypothesis is correct that mTOR inhibition can reduce HCC tumour growth while simultaneously providing immunosuppression to protect the liver allograft from rejection, patients should experience less post-transplant problems with HCC recurrence, and therefore could expect a longer and better quality of life. A positive outcome will likely change the standard of posttransplant immunosuppressive care for LT patients with HCC. Introduction: It has been proposed that individual genetic variation contributes to the course of severe infections and sepsis. Recent studies of single nucleotide polymorphisms (SNPs) within the endotoxin receptor and its signaling system showed an association with the risk of disease development. This study aims to examine the response associated with genetic variations of TLR4, the receptor for bacterial LPS, and a central intracellular signal transducer (TIRAP/Mal) on cytokine release and for susceptibility and course of severe hospital acquired infections in distinct patient populations. Methods: Three intensive care units in tertiary care university hospitals in Greece and Germany participated. 375 and 415 postoperative patients and 159 patients with ventilator associated pneumonia (VAP) were included. TLR4 and TIRAP/Mal polymorphisms in 375 general surgical patients were associated with risk of infection, clinical course and outcome. In two prospective studies, 415 patients following cardiac surgery and 159 patients with newly diagnosed VAP predominantly caused by Gram-negative bacteria were studied for cytokine levels in-vivo and after ex-vivo monocyte stimulation and clinical course. Results: Patients simultaneously carrying polymorphisms in TIRAP/Mal and TLR4 and patients homozygous for the TIRAP/Mal SNP had a significantly higher risk of severe infections after surgery (odds ratio (OR) 5.5; confidence interval (CI): 1.34 - 22.64; P = 0.02 and OR: 7.3; CI: 1.89 - 28.50; P < 0.01 respectively). Additionally we found significantly lower circulating cytokine levels in double-mutant individuals with ventilator associated pneumonia and reduced cytokine production in an ex-vivo monocyte stimulation assay, but this difference was not apparent in TIRAP/Mal-homozygous patients. In cardiac surgery patients without infection, the cytokine release profiles were not changed when comparing different genotypes. Conclusions: Carriers of mutations in sequential components of the TLR signaling system may have an increased risk for severe infections. Patients with this genotype showed a decrease in cytokine release when infected which was not apparent in patients with sterile inflammation following cardiac surgery.

Blood vessels form de novo through the tightly regulated programs of vasculogenesis and angiogenesis. Both processes are distinct but one of the steps they share is the formation of a central lumen, when groups of cells organized as vascular cords undergo complex changes to achieve a tube-like morphology. Recently, a protein termed epidermal growth factor-like domain 7 (EGFL7) was described as a novel endothelial cell-derived factor involved in the regulation of the spatial arrangement of cells during vascular tube assembly. With its impact on tubulogenesis and vessel shape EGFL7 joined the large family of molecules governing blood vessel formation. Only recently, the molecular mechanisms underlying EGFL7's effects have been started to be elucidated and shaping of the extracellular matrix (ECM) as well as Notch signaling might very well play a role in mediating its biological effects. Further, findings in knock-out animal models suggest miR-126, a miRNA located within the egfl7 gene, has a major role in vessel development by promoting VEGF signaling, angiogenesis and vascular integrity. This review summarizes our current knowledge on EGFL7 and miR-126 and we will discuss the implications of both bioactive molecules for the formation of blood vessels.
Background: Up to the 1950s, there was an ongoing debate about the diversity of hereditary optic neuropathies, in particular as to whether all inherited optic atrophies can be ascribed to Leber's hereditary optic neuropathy (LHON) or represent different disease entities. In 1954 W. Jaeger published a detailed clinical and genealogical investigation of a large family with explicit autosomal dominant segregation of optic atrophy thus proving the existence of a discrete disease different from LHON, which is nowadays known as autosomal dominant optic atrophy (ADOA). Since the year 2000 ADOA is associated with genomic mutations in the OPA1 gene, which codes for a protein that is imported into mitochondria where it is required for mitochondrial fusion. Interestingly enough, the underlying mutation in this family has not been identified since then. Results: We have reinvestigated this family with the aim to identify the mutation and to further clarify the underlying pathomechanism. Patients showed a classical non-syndromic ADOA. The long term deterioration in vision in the two teenagers examined 50 years later is of particular note 5/20 to 6/120. Multiplex ligation probe amplification revealed a duplication of the OPA1 exons 7-9 which was confirmed by long distance PCR and cDNA analysis, resulting in an in-frame duplication of 102 amino acids. Segregation was verified in 53 available members of the updated pedigree and a penetrance of 88% was calculated. Fibroblast cultures from skin biopsies were established to assess the mitochondrial network integrity and to qualitatively and quantitatively study the consequences of the mutation on transcript and protein level. Fibroblast cultures demonstrated a fragmented mitochondrial network. Processing of the OPA1 protein was altered. There was no correlation of the OPA1 transcript levels and the OPA1 protein levels in the fibroblasts. Intriguingly an overall decrease of mitochondrial proteins was observed in patients' fibroblasts, while the OPA1 transcript levels were elevated. Conclusions: The thorough study of this family provides a detailed clinical picture accompanied by a molecular investigation of patients' fibroblasts. Our data show a classic OPA1-associated non-syndromic ADOA segregating in this family. Cell biological findings suggest that OPA1 is regulated by post-translational mechanisms and we would like to hypothesize that loss of OPA1 function might lead to impaired mitochondrial quality control. With the clinical, genetic and cell biological characterisation of a family described already more than 50 years ago, we span more than half a century of research in optic neuropathies.
Background: Vaccinia virus strain Lister Elstree (VACV) is a test virus in the DVV/RKI guidelines as representative of the stable enveloped viruses. Since the potential risk of laboratory-acquired infections with VACV persists and since the adverse effects of vaccination with VACV are described, the replacement of VACV by the modified vaccinia Ankara strain (MVA) was studied by testing the activity of different chemical biocides in three German laboratories. Methods: The inactivating properties of different chemical biocides (peracetic acid, aldehydes and alcohols) were tested in a quantitative suspension test according to the DVV/RKI guideline. All tests were performed with a protein load of 10% fetal calf serum with both viruses in parallel using different concentrations and contact times. Residual virus was determined by endpoint dilution method. Results: The chemical biocides exhibited similar virucidal activity against VACV and MVA. In three cases intra-laboratory differences were determined between VACV and MVA - 40% (v/v) ethanol and 30% (v/v) isopropanol are more active against MVA, whereas MVA seems more stable than VACV when testing with 0.05% glutardialdehyde. Test accuracy across the three participating laboratories was high. Remarkably inter-laboratory differences in the reduction factor were only observed in two cases. Conclusions: Our data provide valuable information for the replacement of VACV by MVA for testing chemical biocides and disinfectants. Because MVA does not replicate in humans this would eliminate the potential risk of inadvertent inoculation with vaccinia virus and disease in non-vaccinated laboratory workers.
Introduction: Amniotic fluid harbors cells indicative of all three germ layers, and pluripotent fetal amniotic fluid stem cells (AFSs) are considered potentially valuable for applications in cellular therapy and tissue engineering. We investigated whether it is possible to direct the cell fate of AFSs in vivo by transplantation experiments into a particular microenvironment, the mammary fat pad. This microenvironment provides the prerequisites to study stem cell function and the communication between mesenchymal and epithelial cells. On clearance of the endogenous epithelium, the ductal tree can be reconstituted by the transfer of exogenously provided mammary stem cells. Analogously, exogenously provided stem cells from other tissues can be investigated for their potential to contribute to mammary gland regeneration. Methods: We derived pluripotent murine AFSs, measured the expression of stem cell markers, and confirmed their in vitro differentiation potential. AFSs were transplanted into cleared and non cleared fat pads of immunocompromised mice to evaluate their ability to assume particular cell fates under the instructive conditions of the fat-pad microenvironment and the hormonal stimulation during pregnancy. Results: Transplantation of AFSs into cleared fat pads alone or in the presence of exogenous mammary epithelial cells caused their differentiation into stroma and adipocytes and replaced endogenous mesenchymal components surrounding the ducts in co-transplantation experiments. Similarly, transplantation of AFSs into fat pads that had not been previously cleared led to AFS-derived stromal cells surrounding the elongating endogenous ducts. AFSs expressed the marker protein α-SMA, but did not integrate into the myoepithelial cell layer of the ducts in virgin mice. With pregnancy, a small number of AFS-derived cells were present in acinar structures. Conclusions: Our data demonstrate that the microenvironmental cues of the mammary fat pad cause AFSs to participate in mammary gland regeneration by providing mesenchymal components to emerging glandular structures, but do not incorporate or differentiate into ductal epithelial cells.
Poster Presentation from Nineteenth Annual Computational Neuroscience Meeting: CNS*2010 San Antonio, TX, USA. 24-30 July 2010 Statistical models of neural activity are at the core of the field of modern computational neuroscience. The activity of single neurons has been modeled to successfully explain dependencies of neural dynamics to its own spiking history, to external stimuli or other covariates [1]. Recently, there has been a growing interest in modeling spiking activity of a population of simultaneously recorded neurons to study the effects of correlations and functional connectivity on neural information processing (existing models include generalized linear models [2,3] or maximum-entropy approaches [4]). For point-process-based models of single neurons, the time-rescaling theorem has proven to be a useful toolbox to assess goodness-of-fit. In its univariate form, the time-rescaling theorem states that if the conditional intensity function of a point process is known, then its inter-spike intervals can be transformed or “rescaled” so that they are independent and exponentially distributed [5]. However, the theorem in its original form lacks sensitivity to detect even strong dependencies between neurons. Here, we present how the theorem can be extended to be applied to neural population models and we provide a step-by-step procedure to perform the statistical tests. We then apply both the univariate and multivariate tests to simplified toy models, but also to more complicated many-neuron models and to neuronal populations recorded in V1 of awake monkey during natural scenes stimulation. We demonstrate that important features of the population activity can only be detected using the multivariate extension of the test. ...
Poster Presentation from Nineteenth Annual Computational Neuroscience Meeting: CNS*2010 San Antonio, TX, USA. 24-30 July 2010 In order to model extracellular potentials the Line-Source method provides [1] a very powerful and accurate approach. In this method transmembane fluxes are understood as sources for potential distributions which obey the Poission-equation with zero boundary conditions in the infinity. Its solutions reveal that the waveforms are proportional to local transmembrane net currents. The extracellular potentials are comparable small in amplitude and with the aid of their second special derivatives, it is possible to interpret them as additional fluxes to be included into the cable equation having an impact on the membrane potential of surrounding cells [2]. On this basis ephaptic interactions have been studied and have been considered to play a minor role in the network activity. This modeling study provides a new approach based on the first principle of the conservation of charges which leads to a generalized form of the cable equation taking into account the full three-dimensional detail of the cell’s geometry and the presence of the extracellular potential. So instead of coupling the compartment model and the model for extracellular potentials by means of the transmembrane currents, a non-linear system of partial differential equations is solved. Because the abstraction of deviding the cell’s geometry into compartments falls apart, it is possible to examine the contribution of the precise cell geometry to the signal processing while not neglecting the impact which could result from the extracellular potential. Some simulations of propagating action potentials on ramified geometries are going to be shown as well as the resulting distributions of extracellular action potentials.
'One Plus One' entstand während einer über mehrere Monate verteilten Aufnahmestudio-Session der Rolling Stones im Juni und August 1968 in den Olympic Studios in London. Die Band kam ohne Vorbereitung ins Studio und entwickelte den Song 'Sympathy for the Devil' improvisatorisch im Laufe der Sessions. Die Arbeit am Stück begann mit einem Liedtext von Sänger Mick Jagger unter dem Arbeitstitel 'The Devil Is My Name'. Jagger hatte diesen Text unter dem Eindruck des Romans 'Der Meister und Margarita' des russischen Schriftstellers Michail Bulgakov verfasst - ein Buch, das er von seiner Partnerin Marianne Faithfull erhalten hatte. In Bulgakovs Roman stattet der Teufel dem Moskau der 1930er Jahre einen Besuch ab, in dessen Verlauf er viele Menschen tötet oder in den Wahnsinn treibt. Der Film zeigt die Arbeit an dem Song, der auf einem treibenden Samba-Rhythmus aufruht, in den verschiedenen Entwicklungsstufen, in immer neuen Ansätzen, sich dem späteren Format annähernd. Der Song wurde 1968 auf dem Studioalbum 'Beggars Banquet' als erstes von zehn Liedern veröffentlicht.
Bereits seit dem 12. Jahrhundert ist die Politik Irlands geprägt durch Konflikte, rohe Gewalt und Protest, was schließlich in der Teilung des Landes in Nordirland und Irland 1921 endete. Besonders zwischen den 1970er und 1990er Jahren kam es immer wieder zu gewaltsamen Eskalationen, die sich in Form von blutigen Unruhen und Aufständen niederschlugen. Viele irische Musiker griffen seit jeher diese Probleme in ihren Songs auf. So verwundert es nicht, dass sich auch die populärste Rockband der grünen Insel - U2 - nicht nur in ihrem Welthit Sunday, Bloody Sunday mit den Geschehnissen rund um den Blutsonntag auseinandersetzt. Vor allem Frontmann Bono Vox engagiert sich auch außerhalb der Band politisch und sozial. Als Texter des Quartetts beschäftigt er sich in den oftmals gesellschaftskritischen Songs auch mit popkulturell fernen Themen wie Gewalt, Armut und Hungersnot, aber auch der Hoffnung und dem Glauben an Gott.
Glam-Rock war eine der auffälligsten stilistischen Ausprägungen der Popkultur anfangs der 1970er Jahre. Er ist optisch vor allem durch schrille, glitzernde und oft feminine Kostüme und Bühnendarstellungen gekennzeichnet. Das Spiel mit Extravaganz und Androgynität geht aber viel weiter und mündet in ein ironisches Thematisieren von damals als prototypisch geltenden Auftretensstile, Geschlechterrollen und Konventionen der Konzertkultur. Insbesondere stellten sich die Stars in übertreibenden Rollen dar, die gelegentlich in eigens für die Aufführung konzipierten Kunstfiguren zugespitzt wurden. Extrovertiert, mit unkonventionellen Kostümen, reichlich Make-Up und einem starken Maß sexueller Mehrdeutigkeit schockierten die Künstler ihr Publikum und überschritten jegliche traditionelle Vorstellungen vom typischen Verhalten, Aussehen und musikalischem Ausdruck von Akteuren der Rock- und Popmusik.
"Ich glaube nämlich - sagen Sie das nicht weiter - daß es einen echten Documentary Film überhaupt nicht gibt", schrieb Erwin Panofsky im August 1942 an seinen Mit-Immigranten Siegfried Kracauer (Kracauer/Panofsky 1996, 11). Obgleich Michael Wadleighs "Woodstock" noch 28 Jahre nach diesem Argwohn den "Oscar" als best documentary erhielt, illustriert der Film die These Panofskys wie kaum ein anderer.
Im Mai 1969 veröffentlichten The Who ihr Album Tommy. Zum ersten Mal in der Rockgeschichte erzählten die 24 Lieder auf vier Plattenseiten eine einzige Geschichte über ein taubes, stummes und blindes Kind, das zum spirituellen Guru wurde. Sofort bekam das Werk, das mit einem "Libretto" verkauft wurde, den pompösen Namen Rock Opera (auch wenn es eher die Bezeichnung Kantate und nicht Oper verdient hätte); kurz nach seiner Veröffentlichung begann die Band, die Lieder des "Tommy" gerufenen Kindes live in den Vereinigten Staaten und England zu präsentieren. Die ersten Aufnahmen von Teilen von Tommy auf einer Bühne datieren aus diesem Sommer, als The Who die Chance hatten, ihre Show auf dem Woodstock-Festival vorzustellen. Ein Jahr später, zum Zeitpunkt der Aufzeichnung des Konzerts auf der Isle of Wight, war die Band somit bereits fast anderthalb Jahre unterwegs auf Tour und hatte sich zu einer der überzeugendsten Live-Acts der Rockgeschichte entwickelt. 1969 hatte die Band schon einmal auf dem Isle-of-Wight-Festival vor damals 100.000 Zuschauern gespielt. Als The Who die Bühne am 30. August 1970 um zwei Uhr morgens betraten, wartete eine 600.000-köpfige Menschenmenge auf sie.
"I wanted to capture the sights, the sounds, the smells, of a hard-working rock band on the road. And I got that. But I got more, a lot more." Mit diesen Worten leitet Regisseur Rob Reiner, der später für Filme wie "Harry und Sally" und die oscarprämierte Stephen-King-Verfilmung "Misery" verantwortlich war, in seiner Rolle als Dokumentarfilmer Marty DiBergi "This Is Spinal Tap" ein. Der Film ist eine so genannte Mockumentary, eine komödiantisch-parodistische Pseudo-Dokumentation, über die fiktive Metalband Spinal Tap.
Die 1970er Jahre galten als Zeit der Katastrophenfilme. Daran angelehnt bewarb die offizielle Pressemappe "The Kids Are Alright" als "the world's first rock'n'roll disaster movie". Wenn man das Zerschlagen der Gitarren auf offener Bühne (smashing) - The Who waren bekannt dafür, dass besonders Townshend und Moon auf der Bühne regelmäßig ihre Instrumente zerschlugen - denn überhaupt als "Katastrophe" ansehen will (der Film enthält einige derartige Szenen), so werden doch keine Hochhäuser, Passagierschiffe oder ganze Städte zerstört wie in den Filmen des Genres, sondern nur die Bedingungen des Konzerts selbst.
In "The Filth and the Fury" porträtiert Regisseur Julien Temple den Aufstieg und Fall der berühmtberüchtigten Punkrock-Band The Sex Pistols Mitte der 1970er Jahre. Temple hatte zuvor bereits bei "The Great Rock'n'Roll Swindle" Regie geführt. Diesem ersten Werk wurde allerdings immer wieder eine einseitige Schilderung der Ereignisse aus Sicht des ehemaligen Managers der Sex Pistols - Malcom McLaren - vorgeworfen. McLaren soll viele Skandale der Band bewusst forciert haben, um aus der negativen Publicity Profit zu schlagen. Die Idee hinter "The Filth and the Fury" war, die "wahre Bandgeschichte" ans Licht zu bringen und auf Zelluloid zu bannen: "For 20 years our life has been rewritten - wrongly and very badly. It becomes intolerable. At some point you have to say: 'Stop. Here is the real deal.' I've told the truth constantly, but now it's on film. Somehow celluloid adds a touch of realism for most Americans".

Die Aufzeichnungen zu "Stop Making Sense" fanden an drei Abenden im Dezember 1983 im Pantages Theatre in Hollywood statt. David Byrne und seine Band machten dort im Rahmen der Promotiontour zu ihrem damals neuem Album 'Speaking in Tounges' Station. Als Regisseur holten sich die Talking Heads den filmerfahrenen Jonathan Demme, der später auch Blockbuster wie "The Silence of the Lambs" und "Philadelphia" schuf, und den Kameramann Jordan Cronenweth (Blade Runner) in die Tourneecrew. "Stop Making Sense" ist Demmes Dokumentarfilm-Debüt.
'Sting - Bring on the night' bietet sich selbst als Dokumentation und Inszenierung zweier Geburtsstunden an: Zum einen hält der Film das erste Solo-Konzert Stings nach seinem Ausscheiden aus der Gruppe Police fest; zum anderen erblickt Jake, Stings viertes Kind, im März 1985 das Licht der Welt. Gemeinsam haben Vater und Sohn dabei eines: Beiden gelingt der gesunde Start in ein neues Leben. Die Parallele der "Geburten" wirkt gesucht, täuscht eine Naivität vor, die weder Film noch Akteure haben. Die Handlung des Films ist schnell erzählt. Er ist in zwei Abschnitte aufgeteilt: zunächst die heiße Phase der Proben vor dem ersten Solo-Konzert Stings, den zweiten Teil bildet das Konzert selbst. Dass der Abschnitt der Proben genau über neun Tage von der Kameracrew begleitet wird, wird mit kurzen Rückblenden auf die Proben und Entstehung der Songs und Szenen aus dem Kreißsaal unterstrichen - auf die erschließende Metapher hindeutend, neun Tage Konzertvorbereitung in Analogie zu neun Monaten Schwangerschaft zu setzen, die aber durchsichtig ist und sich nur schwer erschließt.
1974 wollten der südafrikanische Trompeter Hugh Masekela und der New Yorker Musiker und Produzent Stewart Levine ein Musikfestival in Afrika organisieren. Sie wandten sich an den Box-Impresario Don King mit der Bitte, das Festival im Vorfeld des Titelkampfes zwischen Muhammad Ali und George Foreman in Kinshasa (in der zentralafrikanischen Republik Zaire) zu veranstalten. Es war von Anfang an geplant, das Festival nach dem Vorbild erfolgreich im Kino ausgewerteter Beispiele - wie etwa dem 'Woodstock'-Film (USA 1970, Michael Wadleigh) - zu dokumentieren. Ein Kamerateam nahm daher das dreitägige Festival, die Vorbereitungen und eine Reihe von Backstage-Szenen auf. Das Kamerateam war prominent besetzt; zu ihm gehörte etwa der bekannte Direct-Cinema-Regisseur Albert Maysles. Auf Grund einer Verletzung von George Foreman musste der Boxkampf um fünf Wochen verschoben werden, was zur Folge hatte, dass das Festival zeitlich getrennt vom Kampf stattfand und dadurch nicht jene internationale Aufmerksamkeit erreichte, die sich die Organisatoren erhofft hatten.
Nach fünfjähriger Tourneepause kehrte David Bowie 1983 mit seiner 'Serious Moonlight World Tour' zurück, die der Veröffentlichung seines kommerziell hoch erfolgreichen Albums 'Let's Dance' folgte. Am 12.9.1983 führte die Tour ihn auch nach Vancouver, Kanada, wo er vor 15.000 Zuschauern im National Exhibition Coliseum auftrat. Der Sender HBO wurde damit beauftragt, einen Mitschnitt des Konzertes zu erstellen, der die besondere Live-Atmosphäre einfangen sollte. Resultat war eine zwanzig Songs umfassende VHS-Kassette, die 1984 erstmals auf den Markt kam und in der neuesten Überarbeitung seit 2006 als offizielle DVD vorliegt.
"You've heard of Oxford Circus, you've heard of Piccadilly Circus, and this is the Rolling Stones Rock and Roll Circus" - als Rolling-Stones-Frontmann Mick Jagger diesen Satz am 11. Dezember 1968 gegen 14 Uhr in der Montur eines Zirkusdirektors in die Kameras der BBC spricht, ahnt er mit Sicherheit noch nicht, was insbesondere er in den folgenden Wochen für einen beispiellosen Zirkus rund um diese Fernsehaufzeichnung veranstalten wird. Zu diesem Zeitpunkt, da die Bänder für die Aufzeichnung des 'Rolling Stones Rock and Roll Circus' gestartet werden, glaubt Jagger wohl noch, dass die TV-Sendung ein entspanntes Aufeinandertreffen mit befreundeten Rock'n'Roll-Größen wird; er glaubt wohl noch, dass der Auftritt der Rolling Stones am Ende des Konzertes zum furiosen Abschluss wird und die Performances der übrigen Bands in den Schatten stellt; und vor allem glaubt er wohl noch, dass der 'Rock and Roll Circus' wenige Wochen nach der Aufzeichnung über die heimischen Fernseher Großbritanniens flimmert. 15 Stunden später denkt Jagger über all dies anders. Aber der Reihe nach.
Geblendet, verwirrt, unbeachtet und trotzdem irgendwie begeistert - so werden sich viele Zuschauer nach dem Besuch des Pink Floyd-Konzertes am 20. Oktober 1994 in der Londoner Earl Court Arena gefühlt haben; und dasselbe Gefühl kommt auf, wenn man sich den daraus entstandenen Konzertmitschnitt P.U.L.S.E ansieht. Genau 30 Jahre nach der Bandgründung ist für Pink Floyd an diesem Abend Technik Trumpf, der Mensch steht in jeder Hinsicht im Hintergrund.
Only the strong survive

Der zum Zeitpunkt des Films 72jährige Don A. Pennebaker und seine Ehefrau Chris Hegedus gelten als Pioniere des amerikanischen Direct Cinema. Mit Filmen wie 'Don't Look Back' (1967) über Bob Dylan oder 'Monterey Pop' (1968) über das gleichnamige Musikfestival an der Westküste der USA haben die beiden sich in der Geschichte des Dokumentarfilms einen Namen gemacht, der weit über die Fans der jeweiligen Musik hinausgeht. Direct Cinema wurde von den beiden als das unmittelbare Abfilmen der Realität verstanden, die sich vor ihrer Kamera befand. Ein Eingreifen in das Geschehen wurde ebenso strikt abgelehnt wie jeglicher Kommentar. Pennebaker schuf damit einerseits Konzertfilme, die auch die Stimmung, das Publikum, das gesamte Setting mit einfingen (wie eben in 'Monterey Pop'). Andererseits gelang ihm etwa in 'Don't Look Back' ein Porträt des jungen Bob Dylan, das über reine Konzertaufnahmen weit hinausgeht und in der Konzentration auf einen Protagonisten auch imstande ist, einiges über das Verhältnis zwischen Fans und Star, deren Projektionen und den Umgang von Akteuren der Rockmusik mit schnell erworbenem Starstatus zu erzählen.

Heavy Metal hat seinen Ursprung im Hard- und Bluesrock der 1960er und 1970er Jahre. Seine musikalischen Vorgänger sind Bands wie Led Zeppelin, Deep Purple und Iron Butterfly. Der Begriff taucht in der Musik erstmals 1968 auf, als Iron Butterfly ihrem Debütalbum den Titel 'Heavy' gaben und Steppenwolf in ihrem Song 'Born to be wild' über "heavy metal thunder" sangen. Die Entstehung des Ausdrucks ist jedoch unklar, sie wird unter anderem Jimi Hendrix zugeordnet, aber auch als Ausdruck für eine neue Musik, die härter ist als Hardrock – "Metall" (= metal) sei eben in der Realität härter als "Stein" (= rock) - angesehen. Als "Geburtsstunde" des Heavy Metal gilt allgemein der 13. Februar 1970. An diesem Tag erschien das selbst betitelte Debütalbum von Black Sabbath, die häufig als erste Metalband der Welt bezeichnet werden und deren Erfolg bis heute anhält.

Als die britische Band 'Radiohead' 1997 den Filmemacher Grant Gee, der zuvor mit seiner Arbeit an U2s 'Zoo-TV Performance' und mit einigen Musikvideos für Aufsehen gesorgt hatte, als Dokumentarfilmer mit auf ihre Welt-Tournee zum Album "Ok Computer" nahm, wusste keiner der Beteiligten, wie diese verlaufen würde. Frühere Tourneen hatten sich allerdings zum Teil als sehr anstrengend für die Band erwiesen. Vor allem der Frontmann Thom Yorke litt immer wieder unter dem Tour-Alltag. Bedenken muss man zusätzlich, dass die Dokumentation nicht als Bericht über eine Erfolgstour geplant werden konnte, weil zu dem Zeitpunkt, an dem Gees Arbeit an "Meeting people is easy" startete, das Album selbst erst zeitgleich zum Film auf den Markt kam.

Die fast zwölf Jahre dazwischen, in denen Musiker wie David Bowie oder Placebo Pixies-Songs neu auflegten und Hollywood den Pixies-Sound für die Filmmusik entdeckte, haben die Band zwar zu einer einflussreichen Indierock-Bands gemacht, ihre vier Mitglieder aber nur kurzzeitig reicher und - jetzt filmisch belegt - keinesfalls reifer werden lassen.
Schon im Jahr 1970 rechnet der noch junge und von Film und Rockmusik gleichermaßen begeisterte Wim Wenders mit einem Genre ab, "das es nicht gibt": mit dem Musikfilm, vor allem mit den "Rock'n'Roll- und Popmusikfilmen" (Wenders 1989, 82), die bis dato entstanden und denen er insgesamt vorwirft, gerade an der Darstellung der Musik zu scheitern. "Sie zeigen mehr ihr Desinteresse, ihr Missfallen oder ihre Verachtung als ihren Gegenstand. Das, was es zu sehen gibt, die Musiker, die Instrumente, die Bühne, die Arbeit, der Spaß oder die Anstrengung, Musik zu machen, erscheint ihnen nicht wert genug, so wie es ist, gezeigt zu werden" (83). Wenders wirft den Musikfilmen vor, nicht dem authentischen Entstehen der Musik im Augenblick, also live auf der Bühne, und nicht dem Ausdruck der Musik gerecht werden, sondern die eigene Sprache der Rockmusik in eine andere, in die des Films zu übertragen.
Der Dokumentationsfilm 'Feel Like Going Home' ist Bestandteil und Herzstück der von Martin Scorsese initiierten und mitproduzierten Miniserie THE BLUES, die sich mit den Hintergründen und Ursprüngen des Blues als musikalisches und soziales Phänomen beschäftigt. Neben Scorsese selbst steuerten die bekannten Regisseure Wim Wenders, Richard Pearce, Charles Burnett, Marc Levin, Mike Figgis und Clint Eastwood eigene Filme zu dieser Reihe bei. Das verbindende Stilmittel der Filmreihe besteht in einem vermeintlich subjektiven Blick auf das Phänomen 'Blues', der auch auf den Zuschauer übertragen werden soll.
Elvis: that's the way it is

Elvis Presley ist sicher eine der eindrücklichsten Verkörperungen des 'American icon'. Von seinen frühen Aufnahmen für Sun Records angefangen wurde er zur Inkarnation einer prototypischen Jugendfigur der 1950er Jahre - seine Person, der oft kopierte Stil des Auftretens, seine erkennbare sexuelle Aggressivität markierten einen bis dahin unbekannten performativen Stil der Musikdarbietung. Presley gehörte unzweifelhaft zu den jugendlichen Rebellen der 1950er wie Marlon Brando und James Dean auch. Insgesamt 31 Filme und öffentliche Auftritte im Fernsehen schlossen sich an die Bilder der Frühzeit an, entwickelten es bis in die späten 1960er weiter; die provokanten Schärfen des Jugendbildes schliffen sich dabei aber ab, die Zeit der weltweiten Jugendunruhen relativierten und domestizierten es.
Die US-amerikanische Alternative Rockband The Eels wurde 1995 durch den Songwriter Mark Oliver Everett gegründet, der der programmatische Kopf der Gruppe geblieben ist. Sie zeichnete sich von Beginn an durch eine ungewöhnliche Experimentierfreude aus, darum bemüht, immer neue unterschiedliche Klangspektren in der Musik der Band zu entfalten. Everett ist der einzige, der noch aus der Erstformation stammt; sämtliche anderen Mitglieder der Gruppe sind z.T. mehrfach ausgetauscht wurden.
Als die USA 2003 die Invasion in den Irak starteten, befürwortete die breite Öffentlichkeit der Staaten den "Krieg gegen den Terror". Der Großteil der amerikanischen Massenmedien wiederholte gebetsmühlenartig die von der Bush-Regierung propagierten Kriegsgründe. Offene Kritik aus den USA war allenfalls von einigen Intellektuellen und Künstlern zu vernehmen. Zu diesen ist auch Neil Young zu zählen, der sich bereits in den 1960ern und 1970ern aktiv für den Frieden engagierte und den Vietnamkrieg scharf verurteilte. Als er damals für kurze Zeit - zwei Alben und einen Auftritt bei dem Woodstock-Festival - bei CSN (Crosby Stills Nash) einstieg und den Bandnamen um das "Y" erweiterte, schrieb er politische Protestsongs, unter anderem den berühmten Titel "Ohio". 2006, so sagt er in seinem Dokumentarfilm "Crosby, Stills, Nash & Young - Déjà vu", sei die Zeit der Protestsongs eigentlich vorbei.
Cream wurde bereits zu ihrer Gründung Mitte der 1960er Jahre von der Musikwelt als die erste "Supergroup" der Rockmusikgeschichte verstanden. Jack Bruce (Bass), Eric Clapton (Gitarre) und Ginger Baker (Schlagzeug), drei renommierte Musiker, die auf langjährige Erfahrungen in anderen Jazz-, Blues- und Rock-Bands zurückblicken konnten, spielten zwischen 1966 und 1968 eine starke Mischung aus Blues, Rock und Psychedelia, die ihnen Anerkennung seitens der Kritiker und hohe Verkaufszahlen sicherte. Trotz wachsender Erfolge fand die Band neben ihrem rapiden Aufstieg jedoch ein sehr schnelles Ende. 1968, das letzte Jahr der Bandexistenz, war stark von Spannungen zwischen ihren Mitgliedern geprägt, vor allem zwischen Bruce und Baker. Nach einer letzten Tournee durch Amerika sollten die Konzerte in der Londoner Royal Albert Hall den längst geplanten Abschied der Band dokumentieren.
Herbert Windt (1894-1965)

Herbert Windt wurde am 15.9.1894 in Senftenberg in der Niederlausitz als Sohn eines Kaufmanns geboren. Seine Familie war sehr musikalisch veranlagt und brachte ihn schon früh zum Klavierspiel und zum Notenstudium. Als junger Mann verließ er die Schule und ging 1910 an das Sternsche Konservatorium, wo er bis zu seiner freiwilligen Meldung zur Teilnahme am Ersten Weltkrieg im Sommer 1914 studierte. 1917 wurde er als Feldwebel bei Verdun so schwer verwundet, dass er zu 75% als kriegsbeschädigt galt und eine Karriere als Dirigent oder Pianist nicht mehr in Frage kam. Windt studierte daher ab 1921 unter dem renommierten Opernkomponisten Franz Schreker an der Hochschule für Musik in Berlin weiter und widmete sich zusehends eigenen Kompositionen, von denen die Oper Andromache (1932) die bedeutendste ist (obgleich sie nur vier Aufführungen erlebte).
'The Truman Show' bietet jede Menge an großen und ernsten Themen und Lesarten: Kritik am Reality–TV und an den modernen Massenmedien allgemein, sowohl an den Machern wie am Publikum, eine religiöse Allegorie oder eine auf Vater–Sohn–Beziehungen, eine Reflexion über das Verhältnis von Autor und fiktionaler Figur oder eine über die soziale und institutionelle Kontrolle unseres alltäglichen Lebens… Aber es ist auch ein Film über die Freude am Spiel mit filmischen Strukturen, in diesem Falle das Spiel mit den Strukturen filmischer Erzählung resp. mit den Ebenen von Fiktionalität, die jedem fiktionalen Text inhärent sind. Beide Elemente, thematischer Ernst und formales Spiel, sind dicht ineinander verwoben.
Peter Weirs Film 'The Truman Show' (USA 1998, Peter Weir) ist zunächst als klassische Dystopie zu lesen. Beispielhaft wird von einer nicht genau definierten nahen Zukunft erzählt, in der es einem Konzern an Privatpersonen statt möglich ist, ein Kind zu adoptieren und unbehelligt für ein kommerzielles Unternehmen zu missbrauchen: Die Produktion einer ultimativen Reality-Show-Soap-Opera. Der Film erzählt über dieses Motiv von einem unkritischen Medienkonsum, der diese Praxis honorierenden Mehrheit der Fernsehzuschauer und nicht zuletzt von der Macht des Kapitals. Die enorme Produktion wird ausschließlich durch genuines Product-Placement und Merchandising ermöglicht.
Ich möchte mich in der Analyse der Musik in 'The Truman Show' auf das Zusammenspiel von Dramaturgie und Musik konzentrieren. Im Zentrum steht die Frage, welche Informationen - wenn überhaupt - der Film über die Musik vermittelt, an welchen Stellen und in welchen Funktionen Musik nicht bloß als Untermalung und Verstärkung von Emotion eingesetzt wird, sondern um Inhalte des Films zu transportieren. Dabei komme ich nicht umhin, meine eigene Lesart des Films einzubringen. Am Anfang stehen bei einem mehr als zehn Jahre alten Film, der allgemein als 'Medienkritik' interpretiert wird, die Erinnerung an die Jahre, in denen der Film produziert wurde.
Künstlerische Konsequenz hat ihren Preis. Der Theoretiker fordert sie vom Praktiker im Namen stilistischer Reinheit oder medienspezifischer Integrität, Stimmigkeit der kompositorischen Methode oder dramaturgischer Effizienz und schert sich wenig um dessen Sachzwänge vor Ort. Jedoch muß heutzutage ein Film- resp. Medienkomponist - mehr denn je - über Fähigkeiten und Fertigkeiten verfügen, die jenseits solider Kenntnisse des traditionellen Tonsatzes und Kontrapunktes liegen
Insgesamt für 34 Filme (wenige davon mit Fortsetzungen) schrieb Dmitrij Sostakovic begleitende Filmmusik. Er galt als guter Improvisator am Klavier, was ihm zu einer Arbeit im Kinotheater der Stummfilmzeit verhalf, die er benötigte, um seine Familie zu unterstützen und sein Studium am Petrograder Konservatorium zu finanzieren, wo er Klavier, Komposition und Kontrapunkt studierte.
Die landläufige Meinung über Meisels Komposition zu "Berlin - Die Sinfonie der Grossstadt" diskreditiert die Musik vorrangig als illustrativ, die Bildinhalte allein verdoppelnd, klischeehaft und deswegen künstlerisch als nicht sonderlich wertvoll. Diese Einstufung trifft, so pauschal und plakativ wie beispielsweise bei Helga de la Motte-Haber und Hans Emons (1980, 60f) sowie Werner Sudendorf (1984, 20ff) deklariert, nicht zu.
Die Musikindustrie hat mit den Jahren ihre Methoden, Musikstile und Bands maßgeschneidert in den Markt hinein zu produzieren, ja sogar Märkte künstlich zu produzieren, immer weiter verfeinert. Ebenso hat die Kritik an diesen Verfahren, Musikstile wie Moden in schnellem Wechsel aufeinander folgen zu lassen und dadurch Konsumbedürfnisse von Fans immer neu anzufachen, diese als 'synthetisch' verdammt - als stehe alledem ein 'natürliches' Prinzip gegenüber, nach dem sich neue Musikstile organisch aus den historischen und individuellen Erfahrungen von Musikern entwickelten.
Dass die Filmgeschichte des frühen Jazz in weiten Teilen noch Forschungsdesiderat geblieben ist, ist insofern erstaunlich, als die performative Komponente – der Auftritt als solcher, aber genauso die mediale Bannung und Verfügbarmachung des 'unwiederholbaren Moments' - unbestritten als eines der wichtigsten Charakteristika von Jazz gilt. Vorrangiges Medium der Speicherung von Jazzgeschichte sind selbstverständlich die Tonträger. Hinsichtlich der Performance von Jazz - also der körperlichen, Identität vorstellenden oder konstruierenden Präsenz der Musikerin/des Musikers - sind aber gerade Bildmedien und insbesondere Filmkunstwerke aussagekräftige Auskunftgeber (Gabbard 2003). Zu den frühesten tonfilmischen Jazzdokumenten gehören die beiden Jazz-Kurzfilme, die Dudley Murphy im Jahr 1929 für RKO schrieb und drehte: die musical shorts 'St. Louis Blues' mit Bessie Smith (Premiere: New York Ende August 1929) und 'Black and Tan Fantasy' mit Duke Ellington (Premiere: New York 8. Dezember 1929).
Hans-Martin Majewski wurde am 14.1.1911 in Schlawe (Pommern) als Sohn eines Veterinärrats geboren. Er besuchte zunächst das Gymnasium in Schlawe, bevor er bis 1930 seine Schulausbildung auf einer staatlichen Bildungsanstalt in der Stadt Köslin beendete. Danach nahm er in Königsberg das Studium der Medizin auf. 1932 wechselte Majewski nach Leipzig, studierte am dortigen Konservatorium bei Hermann Grabner, Kurt Thomas (Chorleitung), Robert Teichmüller (Klavier), Max Hochkoffler, Bruno Walter (Dirigieren) und Max Ludwig. Seine Schwerpunkte waren Theorie und Kompositionslehre, Dirigenten- und Opernschule.
Anmerkungen zur Geschichte und Praxis der Stummfilmmusik : eine Einführung und Gedankensammlung

Vom Beginn der Filmgeschichte und damit auch der Geschichte der Filmmusik an stand das kommerzielle Interesse durchaus im Vordergrund. Das Bestreben, eine möglichst große Zahl von Adressaten zu erreichen, prägte schon immer die Gestaltung der Filme. Der Wandel der Gesellschaftsschichten, aus denen das Publikum stammte, in den ersten Jahrzehnten der Entwicklung der Filmindustrie – zunächst weniger gebildete Zuschauer auf Jahrmärkten, dann ein weltweites Publikum möglichst aller gesellschaftlichen Klassen – bestimmte in zentralen Punkten die Entwicklung der filmischen Inhalte und Formen und somit auch deren musikalische Begleitung. Die Musikauswahl oder Komposition zu Filmen war und ist dementsprechend von damals bis heute von dem von künstlerischer Leitung und Produktion erwünschten Kundenkreis abhängig und zwischen zwei Extremen angesiedelt: populär und avanciert.
Die Struktur moderner Videoclips ist nicht neu. Schon im 16. Jahrhundert versuchte man mit Instrumenten wie dem optischen Cembalo Optisches und Akustisches zu koppeln. Seit über einem halben Jahrhundert versuchen Künstler folglich Bild und Musik zu vereinen. Besonders in den so genannten Konzept-Videos der elektronischen Musik, bei denen Bild und Ton eine Synthese eingehen und die Musik sichtbar gemacht wird, steht der Satz „Ich sehe Musik“ Pate für die Übersetzung der Musik in Bilder. Neben dem Verschwinden des Interpreten aus dem elektronischen Musik-Video ist die Inszenierung von Körperlichkeit ein Thema der Elektroclips – Körper werden häufig in Rückgriff auf die Bilderwelten des Horror- und Rauschfilms visualisiert: dekonstruiert und deformiert. Dies hängt mit den ideologischen Implikationen der elektronischen Musik zusammen, die sich scheinbar am präzisesten innerhalb dieser Bilderwelten artikulieren lassen. Wie dies geschieht, soll hier anhand von Beispielvideos untersucht werden.

Im schleswig-holsteinischen Dorf Wacken treffen einmal im Jahr für drei Tage mehr als 40.000 Heavy Metal Fans auf weniger als 1.800 Anwohner. Anlass ist das 1990 gegründete Wacken Open Air Festival (W:O:A), nach eigenem Bekunden das größte Metal-Open-Air-Festival der Welt. Dabei müssen kulturelle Differenzen und Konflikte zwischen Dorfbewohnern und Festivalbesuchern vorprogrammiert sein. Diesen Gedanken verfolgte anfangs auch die 1966 geborene koreanische Regisseurin von FULL METAL VILLAGE, Sung-Hyung Cho. Sie beschloss „aus Mangel an Alternativen“ (Cho), einen Dokumentarfilm zu drehen, der den Aufprall beider Welten darstellen sollte. Doch im weiteren Verlauf der Produktion änderte Cho ihr Konzept und es entstand ein Film, der es vermied, die Kontraste zu überspitzen, sondern der sich vielmehr auf die Dokumentation des Lebens der Dorfbewohner konzentriert – die das Festival fast ausnahmslos nicht als Einbruch einer fremden Subkultur wahrgenommen, sondern sich glänzend in die Durchführung und Realität des Festivals integriert haben.
Die Filme Ghostrider, Knowing und Next entfalten auf jeweils unterschiedliche Art und Weise die Problematik, wie vor einer nicht nur unbekannten, sondern auch sehr ernsthaft bedrohlichen Zukunft Entscheidungen zu treffen sind, die die sie angehenden moralischen Subjekte in ihren Handlungsoptionen allererst konstituieren. Die Formung der dargestellten Entscheidungsszenarien wäre nicht denkbar ohne unterschiedliche Zeitlichkeitsmodelle, deren Bedeutung in einer jedes moralische Subjekt betreffenden Möglichkeit liegt, sich gegen die Gegenwart zu wenden. Mögen die Filme als einzelne trivial oder allenfalls unterhaltsam erscheinen, so lässt sich in ihrer Konstellation zeigen, inwiefern jedes Handeln sich nicht so sehr für die Zukunft, sondern an der Gegenwart entscheidet.
A new, two-channel instrument for simultaneous NO3 and N2O5 monitoring was used to make the first comprehensive set of nocturnal NOx measurements (NO, NO2, NO3 and N2O5) at the Taunus Observatory, a rural mountain site (Kleiner Feldberg) in South-western Germany. In May 2008, NO3 and N2O5 mixing ratios were well above the instrumental detection limit (a few ppt) on all nights of the campaign and were characterised by large variability. The concentrations of NO3, N2O5 and NO2 were consistent with the equilibrium constant, K2, defining the rates of formation and thermal dissociation of N2O5. A steady-state lifetime analysis is consistent with the loss of nocturnal NOx being dominated by the reaction of NO3 with volatile organic compounds in this forested region, with N2O5 uptake to aerosols of secondary importance. Analysis of a limited dataset obtained at high relative humidity indicated that the loss of N2O5 by reaction with water vapour is less efficient (>factor 3) than derived using laboratory kinetic data. The fraction of NOx present as NO3 and N2O5 reached ~20% on some nights, with night-time losses of NOx competing with daytime losses.
Fractional release factors of long-lived halogenated organic compounds in the tropical stratosphere

Fractional release factors (FRFs) of organic trace gases are time-independent quantities that influence the calculation of Global Warming Potentials and Ozone Depletion Potentials. We present the first set of vertically resolved FRFs for 15 long-lived halocarbons in the tropical stratosphere up to 34 km altitude. They were calculated from measurements on air samples collected on board balloons and a high altitude aircraft. We compare the derived dependencies of FRFs on the mean stratospheric transit times (the so-called mean ages of air) with similarly derived FRFs originating from measurements at higher latitudes and find significant differences. Moreover a comparison with averaged FRFs currently used by the World Meteorological Organisation revealed the limitations of these measures due to their observed vertical and latitudinal variability. The presented data set could be used to improve future ozone level and climate projections.
We synthesised observations of total particle number (CN) concentration from 36 sites around the world. We found that annual mean CN concentrations are typically 300–2000 cm -3 in the marine boundary layer and free troposphere (FT) and 1000–10 000 cm -3 in the continental boundary layer (BL). Many sites exhibit pronounced seasonality with summer time concentrations a factor of 2–10 greater than wintertime concentrations. We used these CN observations to evaluate primary and secondary sources of particle number in a global aerosol microphysics model. We found that emissions of primary particles can reasonably reproduce the spatial pattern of observed CN concentration (R2=0.46) but fail to explain the observed seasonal cycle (R2=0.1). The modeled CN concentration in the FT was biased low (normalised mean bias, NMB=& -88%) unless a secondary source of particles was included, for example from binary homogeneous nucleation of sulfuric acid and water (NMB= -25%). Simulated CN concentrations in the continental BL were also biased low (NMB= -74%) unless the number emission of anthropogenic primary particles was increased or a mechanism that results in particle formation in the BL was included. We ran a number of simulations where we included an empirical BL nucleation mechanism either using the activation-type mechanism (nucleation rate, J, proportional to gas-phase sulfuric acid concentration to the power one) or kinetic-type mechanism (J proportional to sulfuric acid to the power two) with a range of nucleation coefficients. We found that the seasonal CN cycle observed at continental BL sites was better simulated by BL particle formation (R2=0.3) than by increasing the number emission from primary anthropogenic sources (R2=0.18). The nucleation constants that resulted in best overall match between model and observed CN concentrations were consistent with values derived in previous studies from detailed case studies at individual sites. In our model, kinetic and activation-type nucleation parameterizations gave similar agreement with observed monthly mean CN concentrations.
River flow regimes, including long-term average flows, seasonality, low flows, high flows and other types of flow variability, play an important role for freshwater ecosystems. Thus, climate change affects freshwater ecosystems not only by increased temperatures but also by altered river flow regimes. However, with one exception, transferable quantitative relations between flow alterations and ecological responses have not yet been derived. While discharge decreases are generally considered to be detrimental for ecosystems, the effect of future discharge increases is unclear. As a first step towards a global-scale analysis of climate change impacts on freshwater ecosystems, we quantified the impact of climate change on five ecologically relevant river flow indicators, using the global water model WaterGAP 2.1g to simulate monthly time series of river discharge with a spatial resolution of 0.5 degrees. Four climate change scenarios based on two global climate models and two greenhouse gas emissions scenarios were evaluated. We compared the impact of climate change by the 2050s to the impact of water withdrawals and dams on natural flow regimes that had occurred by 2002. Climate change was computed to alter seasonal flow regimes significantly (i.e. by more than 10%) on 90% of the global land area (excluding Greenland and Antarctica), as compared to only one quarter of the land area that had suffered from significant seasonal flow regime alterations due to dams and water withdrawals. Due to climate change, the timing of the maximum mean monthly river discharge will be shifted by at least one month on one third on the global land area, more often towards earlier months (mainly due to earlier snowmelt). Dams and withdrawals had caused comparable shifts on less than 5% of the land area only. Long-term average annual river discharge is predicted to significantly increase on one half of the land area, and to significantly decrease on one quarter. Dams and withdrawals had led to significant decreases on one sixth of the land area, and nowhere to increases. Thus, by the 2050s, climate change may have impacted ecologically relevant river flow characteristics more strongly than dams and water withdrawals have up to now. The only exception refers to the decrease of the statistical low flow Q90, with significant decreases both by past water withdrawals and future climate change on one quarter of the land area. However, dam impacts are likely underestimated by our study. Considering long-term average river discharge, only a few regions, including Spain, Italy, Iraq, Southern India, Western China, the Australian Murray Darling Basin and the High Plains Aquifer in the USA, all of them with extensive irrigation, are expected to be less affected by climate change than by past anthropogenic flow alterations. In some of these regions, climate change will exacerbate the discharge reductions, while in others climate change provides opportunities for reducing past reductions. Emissions scenario B2 leads to only slightly reduced alterations of river flow regimes as compared to scenario A2 even though emissions are much smaller. The differences in alterations resulting from the two applied climate models are larger than those resulting from the two emissions scenarios. Based on general knowledge about ecosystem responses to flow alterations and data related to flow alterations by dams and water withdrawals, we expect that the computed climate change induced river flow alterations will impact freshwater ecosystems more strongly than past anthropogenic alterations.
This paper investigates the potential impact of secondary information on rainfall mapping applying Ordinary Kriging. Secondary information tested is a natural area indicator, which is a combination of topographic features and weather conditions. Cross validation shows that secondary information only marginally improves the final mapping, indicating that a one-day accumulation time is possibly too short.
Development of a Bioaerosol single particle detector (BIO IN) for the Fast Ice Nucleus CHamber FINCH

In this work we present the setup and first tests of our new BIO IN detector. This detector was constructed to classify atmospheric ice nuclei (IN) for their biological content. It is designed to be coupled to the Fast Ice Nucleus CHamber FINCH. If one particle acts as an ice nucleus, it will be at least partly covered with ice at the end of the development section of the FINCH chamber. The device combines an auto-fluorescence detector and a circular depolarization detector for simultaneous detection of biological material and discrimination between water droplets, ice crystals and non activated large aerosol particles. The excitation of biological material with UV light and analysis of auto-fluorescence is a common principle used for flow cytometry, fluorescence microscopy, spectroscopy and imaging. The detection of auto-fluorescence of airborne single particles demands some more experimental effort. However, expensive commercial sensors are available for special purposes, e.g. size distribution measurements. But these sensors will not fit the specifications needed for the FINCH IN counter (e.g. high sample flow of up 10 LPM). The newly developed -low cost- BIO IN sensor uses a single high-power UV LED for the electronic excitation instead of much more expensive UV lasers. Other key advantages of the new sensor are the low weight, compact size, and the little effect on the aerosol sample, which allows it to be coupled with other instruments for further analysis. The instrument will be flown on one of the first missions of the new German research aircraft "HALO" (High Altitude and LOng range).
Processes occurring in the tropical upper troposphere (UT), the Tropical Transition Layer (TTL), and the lower stratosphere (LS) are of importance for the global climate, for stratospheric dynamics and air chemistry, and for their influence on the global distribution of water vapour, trace gases and aerosols. In this contribution we present aerosol and trace gas (in-situ) measurements from the tropical UT/LS over Southern Brazil, Northern Australia, and West Africa. The instruments were operated on board of the Russian high altitude research aircraft M-55 "Geophysica" and the DLR Falcon-20 during the campaigns TROCCINOX (Araçatuba, Brazil, February 2005), SCOUT-O3 (Darwin, Australia, December 2005), and SCOUT-AMMA (Ouagadougou, Burkina Faso, August 2006). The data cover submicron particle number densities and volatility from the COndensation PArticle counting System (COPAS), as well as relevant trace gases like N2O, ozone, and CO. We use these trace gas measurements to place the aerosol data into a broader atmospheric context. Also a juxtaposition of the submicron particle data with previous measurements over Costa Rica and other tropical locations between 1999 and 2007 (NASA DC-8 and NASA WB-57F) is provided. The submicron particle number densities, as a function of altitude, were found to be remarkably constant in the tropical UT/LS altitude band for the two decades after 1987. Thus, a parameterisation suitable for models can be extracted from these measurements. Compared to the average levels in the period between 1987 and 2007 a slight increase of particle abundances was found for 2005/2006 at altitudes with potential temperatures, theta, above 430 K. The origins of this increase are unknown except for increases measured during SCOUT-AMMA. Here the eruption of the Soufrière Hills volcano in the Caribbean caused elevated particle mixing ratios. The vertical profiles from Northern hemispheric mid-latitudes between 1999 and 2006 also are compact enough to derive a parameterisation. The tropical profiles all show a broad maximum of particle mixing ratios (between theta ~ 340 K and 390 K) which extends from below the TTL to above the thermal tropopause. Thus these particles are a "reservoir" for vertical transport into the stratosphere. The ratio of non-volatile particle number density to total particle number density was also measured by COPAS. The vertical profiles of this ratio have a maximum of 50% above 370 K over Australia and West Africa and a pronounced minimum directly below. Without detailed chemical composition measurements a reason for the increase of non-volatile particle fractions cannot yet be given. However, half of the particles from the tropical "reservoir" contain compounds other than sulphuric acid and water. Correlations of the measured aerosol mixing ratios with N2O and ozone exhibit compact relationships for the tropical data from SCOUT-AMMA, TROCCINOX, and SCOUT-O3. Correlations with CO are more scattered probably because of the connection to different pollution source regions. We provide additional data from the long distance transfer flights to the campaign sites in Brazil, Australia, and West-Africa. These were executed during a time window of 17 months within a period of relative volcanic quiescence. Thus the data represent a "snapshot picture" documenting the status of a significant part of the global UT/LS fine aerosol at low concentration levels 15 years after the last major (i.e., the 1991 Mount Pinatubo) eruption. The corresponding latitudinal distributions of the measured particle number densities are presented in this paper to provide data of the UT/LS background aerosol for modelling purposes.
Abrupt climate changes of the last deglaciation detected in a Western Mediterranean forest record

Abrupt changes in Western Mediterranean climate during the last deglaciation (20 to 6 cal ka BP) are detected in marine core MD95-2043 (Alboran Sea) through the investigation of high-resolution pollen data and pollen-based climate reconstructions by the modern analogue technique (MAT) for annual precipitation (Pann) and mean temperatures of the coldest and warmest months (MTCO and MTWA). Changes in temperate Mediterranean forest development and composition and MAT reconstructions indicate major climatic shifts with parallel temperature and precipitation changes at the onsets of Heinrich stadial 1 (equivalent to the Oldest Dryas), the Bölling-Allerød (BA), and the Younger Dryas (YD). Multi-centennial-scale oscillations in forest development occurred throughout the BA, YD, and early Holocene. Shifts in vegetation composition and (Pann reconstructions indicate that forest declines occurred during dry, and generally cool, episodes centred at 14.0, 13.3, 12.9, 11.8, 10.7, 10.1, 9.2, 8.3 and 7.4 cal ka BP. The forest record also suggests multiple, low-amplitude Preboreal (PB) climate oscillations, and a marked increase in moisture availability for forest development at the end of the PB at 10.6 cal ka BP. Dry atmospheric conditions in the Western Mediterranean occurred in phase with Lateglacial events of high-latitude cooling including GI-1d (Older Dryas), GI-1b (Intra-Allerød Cold Period) and GS-1 (YD), and during Holocene events associated with high-latitude cooling, meltwater pulses and N. Atlantic ice-rafting. A possible climatic mechanism for the recurrence of dry intervals and an opposed regional precipitation pattern with respect to Western-central Europe relates to the dynamics of the westerlies and the prevalence of atmospheric blocking highs. Comparison of radiocarbon and ice-core ages for well-defined climatic transitions in the forest record suggests possible enhancement of marine reservoir ages in the Alboran Sea by 200 years (surface water age 600 years) during the Lateglacial.

Hans Sahl, aus Dresden gebürtig, gehörte zu jenen brillanten Intellektuellen, die bereits im geistigen Leben der Weimarer Republik eine Rolle spielten, vor den Nazis fliehen mußten und als ungeliebte Heimkehrer in der Bundesrepublik lange vergessen blieben. Erst in seinen letzten Lebensjahren, die der fast erblindete Autor in Tübingen verbrachte, wurde er mit Preisen überschüttet, wurden seine Gedichte und Romane neuaufgelegt. “Wir sind die Letzten“, hieß eine Gedichtsammlung, die 1941 erschien, in der sich dieser „Trödler des Unbegreiflichen“, wie er sich nannte, als Moralist und Wahrheitsfanatiker von hohen Graden zu erkennen gab.
“Es ist weder Schwäche noch Minderwertigkeit des Erotischen, wenn es seiner Art nach auf gespanntem Fuß mit der Treue steht, vielmehr bedeutet es an ihm das Abzeichen seines Aufstiegs zu noch weiteren Zusammenhängen. Das Paar Lou/ Nietzsche hat die Phantasie späterer Generationen beflügelt, dieses Zuendedenken und –leben nicht erfüllter Wünsche und Sehnsüchte hat das Kolorit beider Erinnerung wesentlich überzeichnet.
Eines der markantesten Symbole der Advents- und Weihnachtszeit sind die in unterschiedlichsten Farben, Formen und sogar Düften angebotenen Kerzen. Ihr Licht gibt der Jahreszeit eine besondere Atmosphäre, sie unterstützen neben anderen Dekorationsartikeln die erwünschte besinnliche Stimmung. Kerzen stehen traditionell sinnbildlich für Licht, Wärme und Reinheit und symbolisieren damit im Christentum das Lebenslicht des Menschen und Christus als das Licht der Welt. Man könnte nun meinen, dass Kerzen auf den Seiten des Bochumer Botanischen Vereins nichts zu suchen haben, sondern eher dem Bereich Chemie (Stearin) oder Zoologie (Bienenwachs) zugeordnet werden sollten. Aber die Botanik hat auch hier schon immer eine wesentliche Rolle gespielt und steht auch heute noch im wörtlichen Sinne im Zentrum jeder Kerze, denn Wachs brennt ohne sie nicht: die Kerzendochte. Dabei ist die Erfindung des Dochtes weitaus älter als die der Kerzen. Bei den alten Griechen z. B. waren Kerzen noch weitestgehend unbekannt, man verwendete Öllampen, die einen (meist) pflanzlichen Docht enthielten.
Krokusse gehören zu den frühesten Boten des Frühlings. Wenn es im Spätherbst und Winter wärmer ist als gewöhnlich, sprießen schon sehr zeitig die charakteristischen grasartigen, aber fleischigen Blätter mit dem deutlich abgesetzten weißen Mittelstreifen (Unterschied zum Milchstern [Ornithogalum], bei dem das Weiß eher verschwommen und undeutlich abgesetzt ist). Die Blüten der ersten Arten öffnen sich bei uns ab dem zweiten Februardrittel, je nachdem wie die Witterung es zulässt. Diese Schwertliliengewächse (Iridaceae) besitzen wie die meisten Einkeimblättrigen eine einfache Blütenhülle, diese ist trichter- bis becherförmig ("Große Trichterblume"), mit gleich langen Abschnitten und einem röhrig erweiterten Schlund. Die Blüten öffnen sich nur bei Sonnenschein.
Diese Flechtenart gehört zu der umfangreichen Gattung Cladonia, den Becher-, Säulen- und Rentierflechten. Innerhalb dieser Gattung bilden die Rentierflechten eine eigene Gruppe nah verwandter Strauchflechten, die aufrechte, reich verzweigte Stämmchen aufweisen und an ein unbelaubtes Bäumchen oder ein übermäßig gegliedertes Geweih erinnern. Sie werden auch als eigene Gattung Cladina in einigen Bearbeitungen abgetrennt.
Der Bochumer Stadtpark zeichnet sich durch eine große Anzahl von seltenen Gehölzen aus, die in vielen Fällen schon eine bemerkenswerte Größe erreicht haben. Eine von den etwa 100 unterschiedlichen Baumarten des Parks ist der Oregon-Ahorn, der als direkte Übersetzung des lateinischen Namens auch Großblättriger Ahorn genannt wird. Das Exemplar steht im Südwesten des Parks oberhalb des Weges der nördlich des Museumsteiches verläuft. Die Art wird außerhalb von Gehölzsammlungen in Deutschland nicht gepflanzt und ist daher bei uns selten. Der Baum im Stadtpark dürfte mit seiner Größe von ca. 25 m und einem Stammumfang von 2,50 m auch bundesweit zu den Raritäten gehören.
Recently, the first part of the morphological revision of the Southeast Asian water monitor lizards of the Varanus salvator (Laurenti, 1768) species group provided a taxonomic overview over the members of this successful and widespread species complex (Koch et al. 2007). There, the Philippine taxa marmoratus, nuchalis and cumingi were reelevated to species status due to diagnostic morphological characteristics, e.g. significantly enlarged scales on the neck region. In this second part of the ongoing revision, these three species are re-investigated using additional voucher specimens and advanced statistical techniques including canonical variates analysis and principal component analysis. Our new investigations indicate that V. marmoratus represents a composite species, comprising at least three distinct taxa. Hence, the populations of the Sulu Archipelago (Tawi-Tawi Island) and those of the Palawan region are described as new species, viz. Varanus rasmusseni sp. nov. and V. palawanensis sp. nov., respectively. The allopatric island populations of V. cumingi inhabiting Samar, Leyte, and Bohol (the East Visayan subregion) show characteristic and geographically correlated colour patterns distinct from the type locality Mindanao (the second subregion of Greater Mindanao), warranting subspecific partition of this species. The new subspecies is named V. cumingi samarensis ssp. nov. In contrast, the taxonomic status of V. nuchalis remained unchanged, although this species shows some considerable variation in colour pattern. The systematic chapters are supplemented with notes about biology and conservation status. The hitherto underestimated diversity and zoogeography of Philippine water monitors is discussed in the light of Pleistocene sea level fluctuations. Finally, we introduce a scenario for the evolution and spread of Southeast Asian water monitor lizards and provide an identification key for the Philippine members of the V. salvatoI' complex.
Poster presentation at 5th German Conference on Cheminformatics: 23. CIC-Workshop Goslar, Germany. 8-10 November 2009 We demonstrate the theoretical and practical application of modern kernel-based machine learning methods to ligand-based virtual screening by successful prospective screening for novel agonists of the peroxisome proliferator-activated receptor gamma (PPARgamma) [1]. PPARgamma is a nuclear receptor involved in lipid and glucose metabolism, and related to type-2 diabetes and dyslipidemia. Applied methods included a graph kernel designed for molecular similarity analysis [2], kernel principle component analysis [3], multiple kernel learning [4], and, Gaussian process regression [5]. In the machine learning approach to ligand-based virtual screening, one uses the similarity principle [6] to identify potentially active compounds based on their similarity to known reference ligands. Kernel-based machine learning [7] uses the "kernel trick", a systematic approach to the derivation of non-linear versions of linear algorithms like separating hyperplanes and regression. Prerequisites for kernel learning are similarity measures with the mathematical property of positive semidefiniteness (kernels). The iterative similarity optimal assignment graph kernel (ISOAK) [2] is defined directly on the annotated structure graph, and was designed specifically for the comparison of small molecules. In our virtual screening study, its use improved results, e.g., in principle component analysis-based visualization and Gaussian process regression. Following a thorough retrospective validation using a data set of 176 published PPARgamma agonists [8], we screened a vendor library for novel agonists. Subsequent testing of 15 compounds in a cell-based transactivation assay [9] yielded four active compounds. The most interesting hit, a natural product derivative with cyclobutane scaffold, is a full selective PPARgamma agonist (EC50 = 10 ± 0.2 microM, inactive on PPARalpha and PPARbeta/delta at 10 microM). We demonstrate how the interplay of several modern kernel-based machine learning approaches can successfully improve ligand-based virtual screening results.
Poster presentation at 5th German Conference on Cheminformatics: 23. CIC-Workshop Goslar, Germany. 8-10 November 2009 Protein kinases are important targets for drug development. The almost identical protein folding of kinases and the common co-substrate ATP leads to the problem of inhibitor selectivity. Type II inhibitors, targeting the inactive conformation of kinases, occupy a hydrophobic pocket with less conserved surrounding amino acids. Human polo-like kinase 1 (Plk1) represents a promising target for approaches to identify new therapeutic agents. Plk1 belongs to a family of highly conserved serine/threonine kinases, and is a key player in mitosis, where it modulates the spindle checkpoint at metaphase/anaphase transition. Plk1 is over-expressed in all today analyzed human tumors of different origin and serves as a negative prognostic marker in cancer patients. The newly identified inhibitor, SBE13, a vanillin derivative, targets Plk1 in its inactive conformation. This leads to selectivity within the Plk family and towards Aurora A. This selectivity can be explained by docking studies of SBE13 into the binding pocket of homology models of Plk1, Plk2 and Plk3 in their inactive conformation. SBE13 showed anti-proliferative effects in cancer cell lines of different origins with EC50 values between 5 microM and 39 microM and induced apoptosis. Increasing concentrations of SBE13 result in increasing amounts of cells in G2/M phase 13 hours after double thymidin block of HeLa cells. The kinase activity of Plk1 was inhibited with an IC50 of 200 pM. Taken together, we could show that carefully designed structure-based virtual screening is well-suited to identify selective type II kinase inhibitors targeting Plk1 as potential anti-cancer therapeutics.
Influenza A (H1N1) 2009 : impact on Frankfurt in due consideration of health care and public health

Background: In April 2009 a novel influenza A H1N1/2009 virus was identified in Mexico and in the United States which quickly spread around the world. Most of the countries established infection surveillance systems in order to track the number of (laboratory-confirmed) H1N1 cases, hospitalizations and deaths. Methods: The impact of the emergence of the novel pandemic (H1N1) 2009 virus on Frankfurt was statistically evaluated by the Health Protection Authority, City of Frankfurt am Main. Vaccination rates of the health care workers (HCWs) of the University Hospital Frankfurt were measured by the Occupational Health Service. Results: Although the virulence of pandemic (H1N1) 2009 seems to be comparable with seasonal influenza, a major patient load and wave of hospital admissions occurred in the summer of 2009. Even though the 2009 vaccination rate of the University Hospital Frankfurt (seasonal influenza [40.5%], swine flu [36.3%]) is better than the average annual uptake of influenza vaccine in the German health care system (approximately 22% for seasonal and 15% for swine flu), vaccination levels remain insufficient. However, physicians were significantly (p < 0.001) more likely to have been vaccinated against swine flu and seasonal influenza than nurses. Conclusions: The outbreak of the pandemic (H1N1) 2009 in April 2009 provided a major challenge to health services around the world. Nosocomial transmission of H1N1/2009 has been documented. Present experience should be used to improve pandemic preparedness plans and vaccination programs ought to target as many HCWs as possible.
Due to an increasing awareness of the potential hazardousness of air pollutants, new laws, rules and guidelines have recently been implemented globally. In this respect, numerous studies have addressed traffic-related exposure to particulate matter using stationary technology so far. By contrast, only few studies used the advanced technology of mobile exposure analysis. The Mobile Air Quality Study (MAQS) addresses the issue of air pollutant exposure by combining advanced high-granularity spatial-temporal analysis with vehicle-mounted, person-mounted and roadside sensors. The MAQS-platform will be used by international collaborators in order 1) to assess air pollutant exposure in relation to road structure, 2) to assess air pollutant exposure in relation to traffic density, 3) to assess air pollutant exposure in relation to weather conditions, 4) to compare exposure within vehicles between front and back seat (children) positions, and 5) to evaluate "traffic zone"- exposure in relation to non-"traffic zone"-exposure. Primarily, the MAQS-platform will focus on particulate matter. With the establishment of advanced mobile analysis tools, it is planed to extend the analysis to other pollutants including including NO2, SO2, nanoparticles, and ozone.
Background: Hemorrhagic shock/resuscitation is associated with aberrant neutrophil activation and organ failure. This experimental porcine study was done to evaluate the effects of Fas-directed extracorporeal immune therapy with a leukocyte inhibition module (LIM) on hemodynamics, neutrophil tissue infiltration, and tissue damage after hemorrhagic shock/resuscitation. Methods: In a prospective controlled double-armed animal trial 24 Munich Mini Pigs (30.3 +/- 3.3 kg) were rapidly haemorrhaged to reach a mean arterial pressure (MAP) of 35 +/- 5 mmHg, maintained hypotensive for 45 minutes, and then were resuscitated with Ringer's solution to baseline MAP. With beginning of resuscitation 12 pigs underwent extracorporeal immune therapy for 3 hours (LIM group) and 12 pigs were resuscitated according to standard medical care (SMC). Haemodynamics, haematologic, metabolic, and organ specific damage parameters were monitored. Neutrophil infiltration was analyzed histologically after 48 and 72 hours. Lipid peroxidation, and apoptosis were specifically determined in lung, bowel, and liver. Results: In the LIM group, neutrophil counts were reduced versus SMC during extracorporeal immune therapy. After 72 hours, the haemodynamic parameters MAP and cardiac output (CO) were significantly better in the LIM group. Histological analyses showed reduction of shock-related neutrophil tissue infiltration in the LIM group, especially in the lungs. Lower amounts of apoptotic cells and lipid peroxidation were found in organs after LIM treatment. Conclusions: Transient Fas-directed extracorporeal immune therapy may protect from posthemorrhagic neutrophil tissue infiltration and tissue damage.
Das religiöse Verständnis des Osterfestes, auf das häufig in Texten, seltener in den Postkartenbildern angespielt wird, bleibt im Hintergrund. Als Motive dienen u.a. Kinder, Lämmer, Küken und Eier, Frühlingslandschaft und Frühlingsblumen, besonders auch Palmkätzchen. Der Schwerpunkt liegt diesmal auf Motiven mit Lämmern. So wie das Lamm traditionell als Zeichen des Lebens und der Unschuld verstanden wird und sein weißes Fell die innere Reinheit und Frömmigkeit symbolisiert, verweist das Osterlamm (Agnus Dei) in christlicher Sicht auf den Gottessohn, der unschuldig für die Menschen gestorben ist.
Zu Goethes Stellung in der Literatur um 1800 gab es vor allem bezüglich der Frage Klassik oder Romantik bzw. zwischen Klassik und Romantik schon lange Diskussionen, die alle die ästhetische Anarchie der Zeit nicht berücksichtigten. Sie kalkulierten daher nicht ein, dass der Schriftsteller, wie oben anhand der "Lehrjahre" gezeigt, nach dem Abschluss eines Werks einfach eine andere, verschiedene Poetik aufgreifen konnte und der ständige Wandel ein Grundmuster seiner literarischen Laufbahn ist. Im folgenden geht es vor allem darum, die unterschiedlichen ästhetischen Auffassungen zu zeigen, die er in dem sich allmählich verschiebenden Literaturbereich der Zeit einnahm. Und so wenig typisch oder vielmehr ausgesprochen individual und sogar goethe- und geniegemäß seine Haltung auch sein mag, sagt sie doch einiges über die Formen der Disloziierung gegenüber überlieferten, eher festen Verhaltensmustern - solchen eines Epochenstils -, die nun möglich waren und sich ergreifen bzw. nutzen ließen. Schließlich war Goethe insgesamt ein recht fruchtbarer Dichter, dessen Werk allerdings größten Schwankungen unterlag - die Spannbreite zwischen dem Besten und dem Belanglosesten (höfische Gebrauchsliteratur, Singspieltexte), ist nirgendwo so groß wie bei ihm. (Bei Tieck zeugen auch die schwächsten Werke noch von einem gewissen Naturtalent und später von großer Kennerschaft seines Metiers (Professionalität); einen "Faust" oder etwas, das Goethes schönsten Gedichten gleichkäme, hat er freilich nicht geschrieben. Hingegen versteht es Goethe, auch Texte zu verfassen, die fast amateurhaft wirken - seine Singspieltexte sind mit den Libretti von da Ponte wirklich nicht zu vergleichen; warum, ist eines der Rätsel, die man bei ihm findet.)
Es gilt zunächst den Verlauf der Freundschaft zwischen dem Maler Johann Heinrich Wilhelm Tischbein (1751-1829) und Goethe wenigstens grob zu skizzieren (1), dann das Campagna-Gemälde etwas genauer in seinem entstehungsgeschichtlichen Kontext zu betrachten (2) und schließlich die Katze aus dem Sack zu lassen, nämlich das eigentliche Bild des Goetheschen Rom-Erlebnisses, das Gegenbild zum Campagna-Gemälde vorzustellen (3). In einem vierten Schritt müssen die Begründungen für ein solches Vorgehen nachgeliefert werden (4), bis dann am Ende ein kurzer Ausblick in die Zukunft geworfen werden kann (5).
Gibt es irgendeine Berechtigung, die ausgetretenen Pfade der Interpretation erneut zu beschreiten und sich den Liedern aus Goethes Roman Wilhelm Meisters Lehrjahre noch einmal und immer wieder zu nähern? Uferlos erscheint die Liste der Forschungsarbeiten, die sich mit diesen Liedern und/oder der Figur Mignon beschäftigen; gerade Mignon hat in der "Deutungstradition" von Goethes Bildungsroman ihren unbestrittenen Platz. Warum also noch einmal? Ein gewisses Anrecht auf ein wiederholtes und neues Lesen der bekannten Stellen genehmigt Goethes Roman selbst, sind es doch gerade Mignons Lieder, die sich zur mehrfachen und dauerhaften "Wiederholung" bekennen, ja die eine dauernde Wiederholung sogar einfordern. Man hat schon von einem "Wiederholungszwang" gesprochen, der durch diese Lieder vorangetrieben werde. Mignons Lieder arbeiten nicht nur häufig mit der Wiederholungsfigur des Refrains; auch ihr Hörer Wilhelm kann nicht umhin, sich diese Lieder mehrfach "wiederholen" zu lassen (234). Könnte es also sein, dass in dieser Wiederholungsmanie ein Hinweis darauf steckt, wie diese Lieder zu verstehen sind?
Ad fontes. Wir wissen, daß fast alle Werke Thomas Manns ihre langen Wurzeln haben, und kaum ein anderer Roman hat längere als die Geschichte des Doktor Faustus. Sie sind, wenn man so sagen darf, über vierzig Jahre lang, und es versteht sich von selbst, daß sich der Stoff über einen so großen Zeitraum wandelte. Wir kennen den berühmten Dreizeilenplan aus dem Jahre 1901 - so wurde er von Thomas Mann selbst am 17. März 1943 benannt. Aber es war - darauf hat Peter de Mendelssohn in seinem Nachwort zur Frankfurter Ausgabe des Romans hingewiesen - nicht ein Dreizeilenplan, sondern es gab derer zwei. Der erste lautete (in Thomas Manns Notizbuch 7 festgehalten): "Zum Roman. Der syphilitische Künstler nähert sich von Sehnsucht getrieben einem reinen, süßen jungen Mädchen, betreibt die Verlobung mit der Ahnungslosen und erschießt sich dicht vor der Hochzeit". Man kann diesen Plan durchaus als Variation der Verführungsgeschichte Fausts und Gretchens lesen, auch wenn von einem Teufelspakt nicht die Rede ist und nicht Gretchen umkommt, sondern der Künstler, der Opfer einer venerischen Erkrankung ist. Aber das "reine, süße junge Mädchen": unverkennbar eine Gretchen-Gestalt. Der zweite Plan, wohl auf Ende 1904 zu datieren, ist von Mendelssohn als Variante und Fortführung der ersten Niederschrift charakterisiert worden, aber bei genauerem Hinsehen zeigt sich, daß es eigentlich ein ganz anderer Plan ist, in den der erste zwar aufgenommen ist, aber darin ein Fremdkörper bleibt.
In diesem Kapitel werden alle Darstellungen zu diesem Themenkreis aus beiden Romanen gesammelt und interpretiert. Die medizingeschichtlichen Hintergründe sollen als erstes anhand der Figur des "Harfners" erläutert werden. [...] Goethes Beschreibung des Harfners bildet eines der Hauptelemente der Romanhandlung. An mehreren Stellen treten bei dieser Figur seelische Abgründe zutage, deren Ursache erst am Ende der Geschichte sichtbar wird. Einen ersten Hinweis liefert der Harfner, als er seinen Herrn Wilhelm um die Entbindung von seinen Pflichten bittet. Er sagt: "Mein Herr, lassen Sie mir mein schaudervolles Geheimnis, und geben Sie mich los! Die Rache, die mich verfolgt, ist nicht des irdischen Richters; ich gehöre einem unerbittlichen Schicksale."
In der vorliegenden Sammlung sind nicht nur die altbekannten, doch immer wieder neu zu entdeckenden Zitate, nicht nur viele geistgeschliffene Sentenzen, die 'den höchsten Sinn im engsten Raum' zusammenfassen, nicht nur allerlei Lebensweisheiten und -wahrheiten in schlichter, einprägsamer Formulierung enthalten, sondern darüber hinaus alle Äußerungen Goethes über die Frau, die für ihn und auch für seine Epoche bedeutungsvoll und charakteristisch sind. Das Belegmaterial ist den Werken (unter Heranziehung von frühen Fassungen und Entwürfen), den naturwissenschaftlichen Schriften, den amtlichen Schriften, den Tagebüchern, Briefen und Gesprächen entnommen, so daß Goethe hier als Dichter, als Wissenschaftler, als Staatsbeamter und nicht zuletzt als Mensch in seinen vielfältigen, öffentlichen und privaten Lebensbezügen, auch im vertraulichen Umgang zu Wort kommt.
Gloël merkt in der Einleitung zu „Goethes Wetzlarer Zeit“ (1911) an, dass „man zwar über Goethes Umwelt in Frankfurt, Leipzig, Straßburg und Weimar gründlich unterrichtet ist, aber nicht in gleichem Maße über die in Wetzlar [...].“ Daran hat sich nicht viel geändert. Gloël ist trotz wenig aufschlussreicher Quellenangaben immer noch Standardautor. Ebenso verhält es sich mit Goethes Beziehung zu Geheimbünden und Freimaurertum. Zwar wecken sie seit je das Interesse seiner Biographen, jedoch steht auch hier die Weimarer Zeit im Vordergrund.
Objective: To summarise the benefits and harms of treatments for women with gestational diabetes mellitus. Design: Systematic review and meta-analysis of randomised controlled trials. Data sources: Embase, Medline, AMED, BIOSIS, CCMed, CDMS, CDSR, CENTRAL, CINAHL, DARE, HTA, NHS EED, Heclinet, SciSearch, several publishers’ databases, and reference lists of relevant secondary literature up to October 2009. Review methods: Included studies were randomised controlled trials of specific treatment for gestational diabetes compared with usual care or "intensified" compared with "less intensified" specific treatment. Results: Five randomised controlled trials matched the inclusion criteria for specific versus usual treatment. All studies used a two step approach with a 50 g glucose challenge test or screening for risk factors, or both, and a subsequent 75 g or 100 g oral glucose tolerance test. Meta-analyses did not show significant differences for most single end points judged to be of direct clinical importance. In women specifically treated for gestational diabetes, shoulder dystocia was significantly less common (odds ratio 0.40, 95% confidence interval 0.21 to 0.75), and one randomised controlled trial reported a significant reduction of pre-eclampsia (2.5 v 5.5%, P=0.02). For the surrogate end point of large for gestational age infants, the odds ratio was 0.48 (0.38 to 0.62). In the 13 randomised controlled trials of different intensities of specific treatments, meta-analysis showed a significant reduction of shoulder dystocia in women with more intensive treatment (0.31, 0.14 to 0.70). Conclusions: Treatment for gestational diabetes, consisting of treatment to lower blood glucose concentration alone or with special obstetric care, seems to lower the risk for some perinatal complications. Decisions regarding treatment should take into account that the evidence of benefit is derived from trials for which women were selected with a two step strategy (glucose challenge test/screening for risk factors and oral glucose tolerance test).
Aus theoretischen und praktischen Gründen gibt es einerseits kaum eine Möglichkeit, die Rezeption von einzelnen belletristischen Büchern und von einzelnen Spielfilmen sogleich schon als Therapie im Sinne der Psychotherapie zu verstehen, andererseits aber scheint es völlig unerlässlich, Mediennutzungen grundsätzlich in einen sehr starken Zusammenhang mit Emotionen und Emotionsproblemen zu bringen - mit einer unausgesetzt notwendigen Bearbeitung von Gefühlen und Gedanken, und diese fortlaufend erforderliche Bearbeitung von Gefühlen und Gedanken kann man dann in den Gesamtkontext einer unerlässlichen Dauer-Therapie stellen.
Zu meinen Schwerpunkten als Professor für Neuere deutsche Literatur gehört neben der Medien- und Kulturwissenschaft insbesondere mit ihren Theoriebildungen auch die Gegenwartsliteratur. Das hört sich gut an, und es mag wohl sein, dass dieser Schwerpunkt dazu geführt hat, dass ich heute hier in dieser Runde sein darf. Tatsächlich aber verbirgt sich hinter einem solchen Schwerpunkt Gegenwartsliteratur eine immense wissenschaftliche Provokation, die viel mit dem Profil meines Faches, der Literaturwissenschaft und ihrer literaturtheoretischen Grundlegung, zu tun hat, vor allem aber mit der Literaturgeschichte. Gleichzeitig ist diese Provokation auch eng mit meiner eigenen intellektuellen und akademischen Biographie verknüpft. Beide Stränge, der wissenschaftliche und theoretische sowie der biographische, treffen sich in meiner persönlichen Auseinandersetzung mit Gegenwartsliteratur, und davon will ich Ihnen heute kurz erzählen.
Das unter Naturschutz stehende Gebiet des Bövinghauser Bachtals (NSG Oberes Ölbachtal in Bochum, NSG Ölbachtal in Dortmund) wurde floristisch, vegetationskundlich und avifaunistisch untersucht. Insbesondere die Ergebnisse der botanischen Untersuchungen werden hier vorgestellt und zur Analyse herangezogen, um Veränderungen festzustellen, die sich seit ersten Untersuchungen Anfang der 1980er Jahre ergeben haben. Insgesamt wurden 437 Sippen an Höheren Pflanzen festgestellt, 366 davon wurden als dauerhaft ansässige Arten gewertet. 19 Arten stehen in einer der Roten Listen, 5 Arten werden auf der Vorwarnliste Nordrhein-Westfalen geführt. Festgestellt wurde insbesondere ein Rückgang oder sogar der Verlust von Arten, die nährstoffarme und/oder eine niedrige Vegetation benötigen. Pflanzensoziologisch wurden die gebietstypischen und dominierenden Pflanzengesellschaften erfasst. Fünf der gefundenen Pflanzengesellschaften stehen in einer Kategorie der Roten Liste. Zu den regional wichtigsten und auch für die Tierwelt bedeutendsten Biotopen gehören die großflächig ausgebildeten Röhrichtgesellschaften im Haupttal des Bövinghauser Baches, die in Form des Scirpo-Phragmitetums sowie der Großseggenrieder des Caricetum gracilis und des Caricetum acutiformis landesweit gefährdet sind. Auffällig ist die "Verbrennnesselung" der Gesellschaften, die sich insbesondere an Wegrändern und in den Feuchtgebieten bemerkbar macht. Hierauf weist auch das reichliche Auftreten von Gesellschaften hin, die aufgrund der Bevorzugung eutropher Standorte landesweit in Ausbreitung begriffen sind. Diese Befunde weisen auch auf die starke Nährstoffbelastung, insbesondere Stickstoffbelastung, des Gebiets hin. Gründe hierfür sind insbesondere der Eintrag durch die angrenzende Landwirtschaft sowie durch Hunde- und Pferdekot. Die Kartierung des Vogelbestands ergab insgesamt 70 Arten. Unter ihnen wurden 44 Brutvogelarten mit 230 Paaren festgestellt. Von den Vogelarten des Gebietes stehen insgesamt 22 auf der Roten Liste, davon 9 Brutvögel. Das Gebiet bietet durch seine Vielgestaltigkeit und hier insbesondere durch die großflächig ausgebildeten Feuchtgebiete einen in der näheren Umgebung einzigartigen Lebensraum für viele seltene Brutvogelarten. So wurden im Untersuchungsjahr Bruten von Feldschwirl, Schwarzkehlchen, Rohrammer und Teichrohrsänger festgestellt. Insgesamt ist festzustellen, dass die Anzahl der Brutpaare besonders in den Röhricht- und Hochstaudenbereichen vor allem auf Grund der häufigen Störungen durch Spaziergänger und freilaufende Hunde relativ niedrig bleibt. Auch für die Gewässerbrüter bestehen derzeit keine ausreichenden Ruhezonen.
Taxonomische Neukombinationen zur Flora Mittel- und Osteuropas, insbesondere Nordrhein-Westfalens

Im Rahmen der Erarbeitung mehrerer umfassender floristischer Inventare von Regionen Mittel- und Osteuropas wurden umfangreiche phytotaxonomische Studien durchgeführt. Als ein Ergebnis dieser Untersuchungen werden Neu- und Umkombinationen bei Sippen verschiedener Familien in den Art- und Unterartstatus vorgenommen und kommentiert sowie einige Gattungen und infragenerische Taxa neu aufgestellt.
Background: Previously, we showed that glioma pathogenesis related protein (GliPR) is induced in CEM T cells upon HIV-1 infection in vitro. To examine whether GliPR plays a role as HIV dependency factor (HDF), we tested the effect of GliPR suppression by siRNA on HIV-1 replication. Results: Induction of GliPR expression by HIV-1 was confirmed in P4-CCR5 cells. When GliPR was suppressed by siRNA, HIV-1 replication was significantly reduced as measured by HIV-1 transcript levels, HIV-1 p24 protein levels, and HIV-1 LTR-driven reporter gene expression, suggesting that GliPR is a cellular co-factor of HIV-1. Microarray analysis of uninfected HeLa cells following knockdown of GliPR revealed, among a multitude of gene expression alterations, a down-regulation of syndecan-1, syndecan-2, protein kinase C alpha (PRKCA), the catalytic subunit beta of cAMP-dependent protein kinase (PRKACB), nuclear receptor co-activator 3 (NCOA3), and cell surface protein CD59 (protectin), all genes having relevance for HIV-1 pathology. Conclusions: The up-regulation of GliPR by HIV-1 and the early significant inhibition of HIV-1 replication mediated by knockdown of GliPR reveal GliPR as an important HIV-1 dependency factor (HDF), which may be exploited for HIV-1 inhibition.
Background: Autism spectrum disorders (ASDs) are a group of highly heritable neurodevelopmental disorders which are characteristically comprised of impairments in social interaction, communication and restricted interests/behaviours. Several cell adhesion transmembrane leucine-rich repeat (LRR) proteins are highly expressed in the nervous system and are thought to be key regulators of its development. Here we present an association study analysing the roles of four promising candidate genes - LRRTM1 (2p), LRRTM3 (10q), LRRN1 (3p) and LRRN3 (7q) - in order to identify common genetic risk factors underlying ASDs.
Methods: In order to gain a better understanding of how the genetic variation within these four gene regions may influence susceptibility to ASDs, a family-based association study was undertaken in 661 families of European ancestry selected from four different ASD cohorts. In addition, a case-control study was undertaken across the four LRR genes, using logistic regression in probands with ASD of each population against 295 ECACC controls.
Results: Significant results were found for LRRN3 and LRRTM3 (P < 0.005), using both single locus and haplotype approaches. These results were further supported by a case-control analysis, which also highlighted additional SNPs in LRRTM3.
Conclusions: Overall, our findings implicate the neuronal leucine-rich genes LRRN3 and LRRTM3 in ASD susceptibility.
Background: Initiated by a clinical case of critical endotracheal tube (ETT) obstruction, we aimed to determine factors that potentially contribute to the development of endotracheal tube obstruction by its inflated cuff. Prehospital climate and storage conditions were simulated. Methods: Five different disposable ETTs (6.0, 7.0, and 8.0 mm inner diameter) were exposed to ambient outside temperature for 13 months. In addition, every second of these tubes was mechanically stressed by clamping its cuffed end between the covers of a metal emergency case for 10 min. Then, all tubes were heated up to normal body temperature, placed within the cock of a syringe, followed by stepwise inflation of their cuffs to pressures of 3 kPa and >=12 kPa, respectively. The inner lumen of the ETT was checked with the naked eye for any obstruction caused by the external cuff pressure. Results: Neither in tubes that were exposed to ambient temperature (range: -12°C to +44°C) nor in those that were also clamped, visible obstruction by inflated cuffs was detected at any of the two cuff pressure levels. Conclusions: We could not demonstrate a critical obstruction of an ETT by its inflated cuff, neither when the cuff was over-inflated to a pressure of 12 kPa or higher, nor in ETTs that had been exposed to unfavorable storage conditions and significant mechanical stress.
Das Wissen um eine antike Kultur ist nicht selten vom Stand der modernen Forschung geprägt. Auf dem Gebiet der Erforschung des antiken Westbalkan hat dieser keineswegs ein Optimum erreicht. Vielmehr haben der politische Umbruch in Osteuropa sowie die Kriege seit den 1990er Jahren in den Nachfolgestaaten des ehemaligen Jugoslawien ein negatives Licht auf jene Länder und seine Menschen geworfen. Diese Bewertung wird darüber hinaus bis in die heutigen Tage paradoxerweise auch auf die Kulturen der Antike übertragen. Dabei bildet die Region an der Adria mit ihren archäologischen Denkmälern aus verschiedenen Jahrtausenden der Menschheitsgeschichte eine der ältesten Kulturlandschaften Europas. In kaum einem anderen Teil des Kontinents finden sich Natur und Kultur derart eng beieinander. Die in albanischer, serbischer, kroatischer oder slowenischer Sprache verfassten wissenschaftlichen Abhandlungen sowie die von den Touristenrouten häufig abseits gelegenen Kulturgüter sind darüber hinaus in Westeuropa zumeist unbekannt. Aus diesem Grund soll dieser Aufsatz einen Beitrag zum kulturellen „Wiederaufbau“ des am Adriatischen Meer und im Dinarischen Gebirge gelegenen antiken Illyricum leisten. Zunächst wird zur allgemeinen Orientierung der geographische Rahmen abgesteckt. Des weiteren soll die geographische sowie topographische Situation der verschiedenen Kulturlandschaften entlang der Adria sowie des Dinarischen Hinterlandes exemplarisch aufgezeigt werden. Schließlich wird versucht, die sowohl historische als auch archäologische „Lücke“ zwischen Griechenland und Italien für die vorrömische Zeit zu schließen.

Background: Environmental noise is ubiquitous in population growth processes, with a well acknowledged potential to affect populations regardless of their sizes. It therefore deserves consideration in population dynamics modelling. The usual approach to incorporating noise into population dynamical models is to make some model parameter(s) (typically the growth rate, the carrying capacity, or both) stochastic and responsive to environment fluctuations. It is however still unclear whether including noise in one or/and another parameter makes a difference to the model performance. Here we investigated this issue with a focus on model fit and predictive accuracy. To do this, we developed three population dynamical models of the Ricker type with the noise included in the growth rate (Model 1), in the carrying capacity (Model 2), and in both (Model 3). We generated several population time series under each model, and used a Bayesian approach to fit the three models to the simulated data. We then compared the model performances in fitting to the data and in forecasting future observations. Results: When the mean intrinsic growth rate, r, in the data was low, the three models had roughly comparable performances, irrespective of the true model and the level of noise. As r increased, Models 1 performed best on data generated from it, and Model 3 tended to perform best on data generated from either Models 2 or Model 3. Model 2 was uniformly outcompeted by the other two models, regardless of the true model and the level of noise. The correlation between the deviance information criterion (DIC) and the mean square error (MSE) used respectively as measure of fit and predictive accuracy was broadly positive. Conclusion: Our results suggested that the way environmental noise is incorporated into a population dynamical model may profoundly affect its performance. Overall, we found that including noise in one or/and another parameter does not matter as long as the mean intrinsic growth rate, r, is low. As r increased, however, the three models performed differently. Models 1 and 3 broadly outperformed Model 2, the first having the advantage of being simple and more computationally tractable. A comforting result emerging from our analysis is the broad positive correlation between MSEs and DICs, suggesting that the latter may also be informative about the predictive performance of a model.
The human immunodeficiency virus type 1 (HIV-1) coreceptor use and viral evolution were analyzed in blood samples from an HIV-1 infected patient undergoing allogeneic stem cell transplantation (SCT). Coreceptor use was predicted in silico from sequence data obtained from the third variable loop region of the viral envelope gene with two software tools. Viral diversity and evolution was evaluated on the same samples by Bayesian inference and maximum likelihood methods. In addition, phenotypic analysis was done by comparison of viral growth in peripheral blood mononuclear cells and in a CCR5 (R5)-deficient T-cell line which was controlled by a reporter assay confirming viral tropism. In silico coreceptor predictions did not match experimental determinations that showed a consistent R5 tropism. Anti-HIV directed antibodies could be detected before and after the SCT. These preexisting antibodies did not prevent viral rebound after the interruption of antiretroviral therapy during the SCT. Eventually, transplantation and readministration of anti-retroviral drugs lead to sustained increase in CD4 counts and decreased viral load to undetectable levels. Unexpectedly, viral diversity decreased after successful SCT. Our data evidence that only R5-tropic virus was found in the patient before and after transplantation. Therefore, blocking CCR5 receptor during stem cell transplantation might have had beneficial effects and this might apply to more patients undergoing allogeneic stem cell transplantation. Furthermore, we revealed a scenario of HIV-1 dynamic different from the commonly described ones. Analysis of viral evolution shows the decrease of viral diversity even during episodes with bursts in viral load.
