This study explored the organization of the semantic field and the conceptual structure of moving experiences by investigating German-language expressions referring to the emotional state of being moved. We used present and past participles of eight psychological verbs as primes in a free word-association task, as these grammatical forms place their conceptual focus on the eliciting situation and on the felt emotional state, respectively. By applying a taxonomy of basic knowledge types and computing the Cognitive Salience Index, we identified joy and sadness as key emotional ingredients of being moved, and significant life events and art experiences as main elicitors of this emotional state. Metric multidimensional scaling analyses of the semantic field revealed that the core terms designate a cluster of emotional states characterized by low degrees of arousal and slightly positive valence, the latter due to a nearly balanced representation of positive and negative elements in the conceptual structure of being moved.

The ALICE Collaboration at the LHC reports measurement of the inclusive production cross section of electrons from semi-leptonic decays of beauty hadrons with rapidity |y| < 0.8 and transverse momentum 1 < pT < 10 GeV/c, in pp collisions at √s = 2.76 TeV. Electrons not originating from semi-electronic decay of beauty hadrons are suppressed using the impact parameter of the corresponding tracks. The production cross section of beauty decay electrons is compared to the result obtained with an alternative method which uses the distribution of the azimuthal angle between heavy-flavour decay electrons and charged hadrons. Perturbative QCD predictions agree with the measured cross section within the experimental and theoretical uncertainties. The integrated visible cross section, σb→e = 3.47 ± 0.40(stat) +1.12 −1.33(sys) ± 0.07(norm) μb, was extrapolated to full phase space using Fixed Order plus Next-to-Leading Log (FONLL) calculations to obtain the total bb production ¯ cross section, σbb¯ = 130 ± 15.1(stat) +42.1 −49.8(sys) +3.4 −3.1(extr) ± 2.5(norm) ± 4.4(BR) μb.
Ursine bears are a mammalian subfamily that comprises six morphologically and ecologically distinct extant species. Previous phylogenetic analyses of concatenated nuclear genes could not resolve all relationships among bears, and appeared to conflict with the mitochondrial phylogeny. Evolutionary processes such as incomplete lineage sorting and introgression can cause gene tree discordance and complicate phylogenetic inferences, but are not accounted for in phylogenetic analyses of concatenated data. We generated a high-resolution data set of autosomal introns from several individuals per species and of Y-chromosomal markers. Incorporating intraspecific variability in coalescence-based phylogenetic and gene flow estimation approaches, we traced the genealogical history of individual alleles. Considerable heterogeneity among nuclear loci and discordance between nuclear and mitochondrial phylogenies were found. A species tree with divergence time estimates indicated that ursine bears diversified within less than 2 My. Consistent with a complex branching order within a clade of Asian bear species, we identified unidirectional gene flow from Asian black into sloth bears. Moreover, gene flow detected from brown into American black bears can explain the conflicting placement of the American black bear in mitochondrial and nuclear phylogenies. These results highlight that both incomplete lineage sorting and introgression are prominent evolutionary forces even on time scales up to several million years. Complex evolutionary patterns are not adequately captured by strictly bifurcating models, and can only be fully understood when analyzing multiple independently inherited loci in a coalescence framework. Phylogenetic incongruence among gene trees hence needs to be recognized as a biologically meaningful signal.
Em alguns de seus trabalhos Talcott Parsons descreveu o estabelecimento das sociedades modernas como um processo de diferenciação de diversas esferas de reconhecimento recíproco. Neste texto eu uso a teoria social do reconhecimento de Parsons para examinar características de conflitos sociais recentes. Começo expondo a descrição que Parsons fez das lutas por reconhecimento ocorridas nas sociedades altamente industrializadas de sua época. Depois tomo a concepção de Parsons sobre conflitos por reconhecimento normativamente orientados para indicar tendências que levaram a uma gradual erosão das estruturas de pacificação social postuladas por Parsons nas últimas décadas do século 20. Descrevo as consequências iniciais dessa desintegração como uma “barbarização” dos conflitos sociais. Por barbarizado entendo um estágio da sociedade em que as lutas por reconhecimento social escalaram e se tornaram anômicas, uma vez que não podem mais ser resolvidas nas esferas sistêmicas estabelecidas de negociação. Este texto revela a importância do conceito de reconhecimento para a teoria social ao acompanhar Parsons na análise de transformações estruturais que atualmente emergem em resposta a conflitos sociais. Banater Schnitten, Pariser Stangen, Russische Elegante : zur Reichweite siebenbürgischer Kochrezepte

The present paper analyses the gastronomic terms with geographical components, which appear especially in the German cooking books, characteristic for the 19th and 20th century from Transylvania. The linguistic analysis (phonology, morpho-syntax, and semantics) and the pragmatics of these terms help us to reflect the economic, historical and cultural realities of those times; the interethnic interferences from the gastronomic field are as profound as those from the cultural and linguistic field are.

Von den in Deutschland und auch in Nordrhein-Westfalen gefährdeten Pflanzenarten zählen neben den Ackerwildkräutern die Wasserpflanzen zu der Gruppe von Pflanzen, die am stärksten vom Verlust oder der negativen Beeinflussung ihrer primären Lebensräume betroffen sind. Im Jahr 2013 wurde der Gewöhnliche Igelschlauch (Baldellia ranunculoides) vom Förderkreis Sporttauchen zur "Wasserpflanze des Jahres" gewählt. In Deutschland und in Nordrhein-Westfalen zählt die Art heute zu den bedrohten Arten und wird in den entsprechenden Roten Listen als "stark gefährdet" angegeben. Im vorliegenden Beitrag werden Systematik und Morphologie sowie Lebensraum und Verbreitung dargestellt.
Hepatitis C virus (HCV) infection leads to the development of hepatic diseases, as well as extrahepatic disorders such as B-cell non-Hodgkin's lymphoma (B-NHL). To reveal the molecular signalling pathways responsible for HCV-associated B-NHL development, we utilised transgenic (Tg) mice that express the full-length HCV genome specifically in B cells and develop non-Hodgkin type B-cell lymphomas (BCLs). The gene expression profiles in B cells from BCL-developing HCV-Tg mice, from BCL-non-developing HCV-Tg mice, and from BCL-non-developing HCV-negative mice were analysed by genome-wide microarray. In BCLs from HCV-Tg mice, the expression of various genes was modified, and for some genes, expression was influenced by the gender of the animals. Markedly modified genes such as Fos, C3, LTβR, A20, NF-κB and miR-26b in BCLs were further characterised using specific assays. We propose that activation of both canonical and alternative NF-κB signalling pathways and down-regulation of miR-26b contribute to the development of HCV-associated B-NHL.
The historical account of Aztec Emperor Auitztol's introduction of the great-tailed grackle Quiscalus mexicanus into the Valley of Mexico (1486–1502) is significant because it documents human translocation of wild birds in Mexico over 500 years ago, before the Spanish Conquest of that land. In the present paper, which defends the account from writings that dispute it, I first review the evidence of how the account was obtained and show that its many details are consistent with what is known from other sources about both the great-tailed grackle and the Aztecs (Nahuas). I then review and examine all published criticisms of the account and explain in detail why they are wrong. The critics have to date presented no persuasive evidence to support their speculation that the Aztecs confused, or might have confused, a natural invasion for an introduction. In contrast to these critics, Bernardino de Sahagún’s research group in the 1500s presented a highly credible, peer-reviewed historical account that documented Aztec introduction of the great-tailed grackle. The pioneering work of these Renaissance Mexican scholars continues to stand as one of the most important records of invasive alien species introduction in ancient times.

Hauptthema des "Buches Stadt der Engel" von Christa Wolf (1929-2011) ist die problematische Feststellung der Realität in unseren heutigen Tagen. Dieses Buch, ein Erzählrahmen funktioniert, der alle Diskurse im Text stützt: den romanhaften, den historischen, den autobiographischen. Sogar die surrealistische Figur eines Schutzengels wird dem Leser geboten, eine Figur, die der Protagonistin als Ausländerin in einem fremden Land hilft, Schicksalsschläge wie die "Entdeckung" der eigenen Vergangenheit als IM Margarete der Stasi zu überwinden. Absicht dieses Essays ist es zu zeigen, wie alle diese Diskurse im Text zu einem literarischen Gewebe artikuliert werden.
Aurora kinase inhibitors displayed activity in pre-clinical neuroblastoma models. Here, we studied the effects of the pan-aurora kinase inhibitor tozasertib (VX680, MK-0457) and the aurora kinase inhibitor alisertib (MLN8237) that shows some specificity for aurora kinase A over aurora kinase B in a panel of neuroblastoma cell lines with acquired drug resistance. Both compounds displayed anti-neuroblastoma activity in the nanomolar range. The anti-neuroblastoma mechanism included inhibition of aurora kinase signalling as indicated by decreased phosphorylation of the aurora kinase substrate histone H3, cell cycle inhibition in G2/M phase, and induction of apoptosis. The activity of alisertib but not of tozasertib was affected by ABCB1 expression. Aurora kinase inhibitors induced a p53 response and their activity was enhanced in combination with the MDM2 inhibitor and p53 activator nutlin-3 in p53 wild-type cells. In conclusion, aurora kinases are potential drug targets in therapy-refractory neuroblastoma, in particular for the vast majority of p53 wild-type cases.

Caspase-2 represents the most conserved member of the caspase family, which exhibits features of both initiator and effector caspases. Using ribonucleoprotein (RNP)-immunoprecipitation assay, we identified the proapoptotic caspase-2L encoding mRNA as a novel target of the ubiquitous RNA-binding protein HuR in DLD-1 colon carcinoma cells. Unexpectedly, crosslinking-RNP and RNA probe pull-down experiments revealed that HuR binds exclusively to the caspase-2-5' untranslated region (UTR) despite that the 3' UTR of the mRNA bears several adenylate- and uridylate-rich elements representing the prototypical HuR binding sites. By using RNAi-mediated loss-of-function approach, we observed that HuR regulates the mRNA and in turn the protein levels of caspase-2 in a negative manner. Silencing of HuR did not affect the stability of caspase-2 mRNA but resulted in an increased redistribution of caspase-2 transcripts from RNP particles to translational active polysomes implicating that HuR exerts a direct repressive effect on caspase-2 translation. Consistently, in vitro translation of a luciferase reporter gene under the control of an upstream caspase-2-5'UTR was strongly impaired after the addition of recombinant HuR, whereas translation of caspase-2 coding region without the 5'UTR is not affected by HuR confirming the functional role of the caspase-2-5'UTR. Functionally, an elevation in caspase-2 level by HuR knockdown correlated with an increased sensitivity of cells to apoptosis induced by staurosporine- and pore-forming toxins as implicated by their significant accumulation in the sub G1 phase and an increase in caspase-2, -3 and poly ADP-ribose polymerase cleavage, respectively. Importantly, HuR knockdown cells remained insensitive toward STS-induced apoptosis if cells were additionally transfected with caspase-2-specific siRNAs. Collectively, our findings support the hypothesis that HuR by acting as an endogenous inhibitor of caspase-2-driven apoptosis may essentially contribute to the antiapoptotic program of adenocarcinoma cells by HuR.
Background: The phagocytic enzyme myeloperoxidase (MPO) acts as a front-line defender against microorganisms. However, increased MPO levels have been found to be associated with complex and calcified atherosclerotic lesions and incident cardiovascular disease. Therefore, this study aimed to investigate a predictive role of MPO, a biomarker of inflammation and oxidative stress, for total and cardiovascular mortality in patients referred to coronary angiography.
Methods and results: MPO plasma concentrations along with eight MPO polymorphisms were determined in 3036 participants of the Ludwigshafen Risk and Cardiovascular Health study (median follow-up 7.75 years). MPO concentrations were positively associated with age, diabetes, smoking, markers of systemic inflammation (interleukin-6, fibrinogen, C-reactive protein, serum amyloid A) and vascular damage (vascular cellular adhesion molecule-1 and intercellular adhesion molecule-1) but negatively associated with HDL-cholesterol and apolipoprotein A-I. After adjustment for cardiovascular risk factors MPO concentrations in the highest versus the lowest quartile were associated with a 1.34-fold risk (95% CI: 1.09–1.67) for total mortality. In the adjusted model the hazard ratio for cardiovascular mortality in the highest MPO quartile was 1.42 (95% CI: 1.07–1.88). Five MPO polymorphisms were positively associated with MPO concentrations but not with mortality. Using Mendelian randomization, we did not obtain evidence for a causal association of MPO with either total or cardiovascular mortality.
Conclusions: MPO concentrations but not genetic variants at the MPO locus are independently associated with risk for total and cardiovascular mortality in coronary artery disease patients.
Association of autoimmune Addison's disease with alleles of STAT4 and GATA3 in European cohorts

Background: Gene variants known to contribute to Autoimmune Addison's disease (AAD) susceptibility include those at the MHC, MICA, CIITA, CTLA4, PTPN22, CYP27B1, NLRP-1 and CD274 loci. The majority of the genetic component to disease susceptibility has yet to be accounted for.
Aim: To investigate the role of 19 candidate genes in AAD susceptibility in six European case-control cohorts.
Methods: A sequential association study design was employed with genotyping using Sequenom iPlex technology. In phase one, 85 SNPs in 19 genes were genotyped in UK and Norwegian AAD cohorts (691 AAD, 715 controls). In phase two, 21 SNPs in 11 genes were genotyped in German, Swedish, Italian and Polish cohorts (1264 AAD, 1221 controls). In phase three, to explore association of GATA3 polymorphisms with AAD and to determine if this association extended to other autoimmune conditions, 15 SNPs in GATA3 were studied in UK and Norwegian AAD cohorts, 1195 type 1 diabetes patients from Norway, 650 rheumatoid arthritis patients from New Zealand and in 283 UK Graves' disease patients. Meta-analysis was used to compare genotype frequencies between the participating centres, allowing for heterogeneity.
Results: We report significant association with alleles of two STAT4 markers in AAD cohorts (rs4274624: P = 0.00016; rs10931481: P = 0.0007). In addition, nominal association of AAD with alleles at GATA3 was found in 3 patient cohorts and supported by meta-analysis. Association of AAD with CYP27B1 alleles was also confirmed, which replicates previous published data. Finally, nominal association was found at SNPs in both the NF-κB1 and IL23A genes in the UK and Italian cohorts respectively.
Conclusions: Variants in the STAT4 gene, previously associated with other autoimmune conditions, confer susceptibility to AAD. Additionally, we report association of GATA3 variants with AAD: this adds to the recent report of association of GATA3 variants with rheumatoid arthritis.
Background: Various kinase inhibitors are known to be ATP-binding cassette (ABC) transporter substrates and resistance acquisition to kinase inhibitors has been associated to increased ABC transporter expression. Here, we investigated the role of the ABC transporters ABCB1, ABCC1, and ABCG2 during melanoma cell resistance acquisition to the V600-mutant BRAF inhibitors PLX4032 (vemurafenib) and PLX4720. PLX4032 had previously been shown to interfere with ABCB1 and ABCG2. PLX4720 had been demonstrated to interact with ABCB1 but to a lower extent than PLX4032.
Findings: PLX4032 and PLX4720 affected ABCC1- and ABCG2-mediated drug transport in a similar fashion. In a panel of 16 V600E BRAF-mutated melanoma cell lines consisting of four parental cell lines and their sub-lines with acquired resistance to PLX4032, PLX4720, vincristine (cytotoxic ABCB1 and ABCC1 substrate), or mitoxantrone (cytotoxic ABCG2 substrate), we detected enhanced ABC transporter expression in 4/4 cytotoxic ABC transporter substrate-resistant, 3/4 PLX4720-resistant, and 1/4 PLX4032-resistant melanoma cell lines.
Conclusion: PLX4032 has the potential to induce ABC transporter expression but this potential is lower than that of PLX4720 or cytotoxic ABC transporter substrates. Since ABC transporters confer multi-drug resistance, this is of relevance for the design of next-line therapies.
It is common practice to use a 30-year period to derive climatological values, as recommended by the World Meteorological Organization. However this convention relies on important assumptions, of which the validity can be examined by deriving the uncertainty inherent to using a limited time-period for deriving climatological values. In this study a new method, aiming at deriving this uncertainty, has been developed with an application to precipitation for a station in Europe (Westdorpe) and one in Africa (Gulu). The weather generator framework is used to produce synthetic daily precipitation time-series that can also be regarded as alternative climate realizations. The framework consists of an improved Markov model, which shows good performance in reproducing the 5-day precipitation variability. The sub-seasonal, seasonal and the inter-annual signals are introduced in the weather generator framework by including covariates. These covariates are derived from an empirical mode decomposition analysis with an improved stability and significance assessment. Introducing covariates was found to substantially improve the monthly precipitation variability for Gulu. From the weather generator, 1,000 synthetic time-series were produced. The divergence between these time-series demonstrates an uncertainty, inherent to using a 30-year period for mean precipitation, of 11 % for Westdorpe and 15 % for Gulu. The uncertainty for precipitation 10-year return levels was found to be 37 % for both sites.
In this study we show how size-resolved measurements of aerosol particles and cloud condensation nuclei (CCN) can be used to characterize the supersaturation of water vapor in a cloud. The method was developed and applied during the ACRIDICON-Zugspitze campaign (17 September to 4 October 2012) at the high-Alpine research station Schneefernerhaus (German Alps, 2650 m a.s.l.). Number size distributions of total and interstitial aerosol particles were measured with a scanning mobility particle sizer (SMPS), and size-resolved CCN efficiency spectra were recorded with a CCN counter system operated at different supersaturation levels.
During the evolution of a cloud, aerosol particles are exposed to different supersaturation levels. We outline and compare different estimates for the lower and upper bounds (Slow, Shigh) and the average value (Savg) of peak supersaturation encountered by the particles in the cloud. A major advantage of the derivation of Slow and Savg from size-resolved CCN efficiency spectra is that it does not require the specific knowledge or assumptions about aerosol hygroscopicity that are needed to derive estimates of Slow, Shigh, and Savg from aerosol size distribution data. For the investigated cloud event, we derived Slow ≈ 0.07–0.25%, Shigh ≈ 0.86–1.31% and Savg ≈ 0.42–0.68%.
Antigen presentation to cytotoxic T lymphocytes via major histocompatibility complex class I (MHC I) molecules depends on the heterodimeric transporter associated with antigen processing (TAP). For efficient antigen supply to MHC I molecules in the ER, TAP assembles a macromolecular peptide-loading complex (PLC) by recruiting tapasin. In evolution, TAP appeared together with effector cells of adaptive immunity at the transition from jawless to jawed vertebrates and diversified further within the jawed vertebrates. Here, we compared TAP function and interaction with tapasin of a range of species within two classes of jawed vertebrates. We found that avian and mammalian TAP1 and TAP2 form heterodimeric complexes across taxa. Moreover, the extra N-terminal domain TMD0 of mammalian TAP1 and TAP2 as well as avian TAP2 recruits tapasin. Strikingly, however, only TAP1 and TAP2 from the same taxon can form a functional heterodimeric translocation complex. These data demonstrate that the dimerization interface between TAP1 and TAP2 and the tapasin docking sites for PLC assembly are conserved in evolution, whereas elements of antigen translocation diverged later in evolution and are thus taxon specific.
This article deals with the topic of dyslexia in adults and its consequences for learning German during university studies; the author explores the extent to which an optimized German text influences understanding. The results of research on dyslexia and psycholinguistic research on text understanding show that depending on the text structure and the simplicity of the linguistic means used, different levels of text understanding may be expected. The main tool of the research is text optimization based on a practical application of the "Hamburg concept of understandability", involving four text properties which make the text understandable to different extents.
Der Beitrag widmet sich einem ausgewählten Bereich öffentlicher Pressesprache, wobei strukturelle, semantische und pragmatische Besonderheiten der sogenannten hölzernen Sprache in der deutschsprachigen Zeitung Neuer Weg (NW) aus Rumänien exemplarisch am Beispiel von Beileidstelegrammen zum Ableben der kommunistischen Führer Josef Wissarionowitsch Stalin und Gheorghe Gheorghiu-Dej (1953 bzw.1965) erfasst werden sollen. Mit der Darstellung von Formulierungspräferenzen in einer ausgewählten Textsorte der öffentlichen schriftlichen Kommunikationspraxis und der Analyse dieses soziokulturell geprägten Sprachgebrauchs hinaus sollen auch die sozial-historischen Bedingungen, die für das Auftauchen der Ausdrucksformen der hölzernen Sprache förderlich waren, eruiert werden.
Neste artigo, pretende-se apontar para uma tensão dialética entre o racional e o irracional, entre o conceito e o não conceitual, atualizando o sentido do não idêntico. Essa atualização configura-se na necessidade de potencializar uma filosofia da educação crítico-formativa com base na perspectivada “expressividade estética”. A questão que se coloca é a seguinte: como podemos compreender o sentido do não idêntico, a partir das necessidades de uma educação crítico-formativa e das condições para efetivá-la no âmbito das reflexões filosófico-educacionais? Desse modo, faz-se uma breve incursão no que consiste o papel da dialética negativa do filósofo Theodor W. Adorno para se pensar o sentido da “razão” (ou das razões) na perspectiva do “não idêntico”. Partindo disso, busca--se apontar as possibilidades da crítica imanente adorniana, cujo escopo manifesta na dialética do sofrimento no âmbito do capitalismo. Bestände, die von Calamagrostis epigejos dominiert werden, gehören zu den charakteristischen Elementen der Vegetation im östlichen Mitteleuropa. In der Literatur wurden Calamagrostis epigejos-Gesellschaften bisher meist als monoton, artenarm und wenig divers beschrieben. Für den Berliner Raum wurden 54 Vegetationsaufnahmen von Brachflächen und ehemaligen Rieselfeldern ausgewertet, in denen C. epigejos dominant war (Deckung >25 %). Die C. epigejos-Dominanzbestände konnten hauptsächlich zwei Gesellschaften zugeordnet werden: der Calamagrostis epigejos-(Galio-Urticetea)-Gesellschaft und der Calamagrostis epigejos-(Onopordetalia/Agropyretalia)-Gesellschaft. Die Calamagrostis epigejos-(Galio-Urticetea)-Gesellschaft wächst auf nährstoffreichen, frischen bis nassen, zum Teil auch zeitweilig überstauten Flächen. Derartige Standortverhältnisse sind vor allem auf eutrophierten ehemaligen Rieselfeldern zu finden. Die Bestände der Calamagrostis epigejos-(Onopordetalia/Agropyretalia)-Gesellschaft kommen auf nährstoffärmeren und trockeneren Standorten vor. Für die mittleren F- und N-Zeigerwerte ergeben sich jeweils signifikante Unterschiede von 1,4 Stufen zwischen beiden Gesellschaften; für die mittleren R-Zeigerwerte beträgt der Unterschied 0,4 Stufen.
Background: Acute leukemia in early age (EAL) is characterized by acquired genetic alterations such as MLL rearrangements (MLL-r). The aim of this case-controlled study was to investigate whether single nucleotide polymorphisms (SNPs) of IKZF1, ARID5B, and CEBPE could be related to the onset of EAL cases (<24 months-old at diagnosis).
Methods: The SNPs (IKZF1 rs11978267, ARID5B rs10821936 and rs10994982, CEBPE rs2239633) were genotyped in 265 cases [169 acute lymphoblastic leukemia (ALL) and 96 acute myeloid leukaemia (AML)] and 505 controls by Taqman allelic discrimination assay. Logistic regression was used to evaluate the association between SNPs of cases and controls, adjusted on skin color and/or age. The risk was determined by calculating odds ratios (ORs) with 95% confidence interval (CI).
Results: Children with the IKZF1 SNP had an increased risk of developing MLL-germline ALL in white children. The heterozygous/mutant genotype in ARID5B rs10994982 significantly increased the risk for MLL-germline leukemia in white and non-white children (OR 2.60, 95% CI: 1.09-6.18 and OR 3.55, 95% CI: 1.57-8.68, respectively). The heterozygous genotype in ARID5B rs10821936 increased the risk for MLL-r leukemia in both white and non-white (OR 2.06, 95% CI: 1.12-3.79 and OR 2.36, 95% CI: 1.09-5.10, respectively). Furthermore, ARID5B rs10821936 conferred increased risk for MLL-MLLT3 positive cases (OR 7.10, 95% CI:1.54-32.68). Our data do not show evidence that CEBPE rs2239633 confers increased genetic susceptibility to EAL.
Conclusions: IKZF1 and CEBPE variants seem to play a minor role in genetic susceptibility to EAL, while ARID5B rs10821936 increased the risk of MLL-MLLT3. This result shows that genetic susceptibility could be associated with the differences regarding MLL breakpoints and partner genes.
Many studies about endocrine pollution in the aquatic environment reveal changes in the reproduction system of biota. We analysed endocrine activities in two rivers in Southern Germany using three approaches: (1) chemical analyses, (2) in vitro bioassays, and (3) in vivo investigations in fish and snails. Chemical analyses were based on gas chromatography coupled with mass spectrometry. For in vitro analyses of endocrine potentials in water, sediment, and waste water samples, we used the E-screen assay (human breast cancer cells MCF-7) and reporter gene assays (human cell line HeLa-9903 and MDA-kb2). In addition, we performed reproduction tests with the freshwater mudsnail Potamopyrgus antipodarum to analyse water and sediment samples. We exposed juvenile brown trout (Salmo trutta f. fario) to water downstream of a wastewater outfall (Schussen River) or to water from a reference site (Argen River) to investigate the vitellogenin production. Furthermore, two feral fish species, chub (Leuciscus cephalus) and spirlin (Alburnoides bipunctatus), were caught in both rivers to determine their gonadal maturity and the gonadosomatic index. Chemical analyses provided only little information about endocrine active substances, whereas the in vitro assays revealed endocrine potentials in most of the samples. In addition to endocrine potentials, we also observed toxic potentials (E-screen/reproduction test) in waste water samples, which could interfere with and camouflage endocrine effects. The results of our in vivo tests were mostly in line with the results of the in vitro assays and revealed a consistent reproduction-disrupting (reproduction tests) and an occasional endocrine action (vitellogenin levels) in both investigated rivers, with more pronounced effects for the Schussen river (e.g. a lower gonadosomatic index). We were able to show that biological in vitro assays for endocrine potentials in natural stream water reasonably reflect reproduction and endocrine disruption observed in snails and field-exposed fish, respectively.
Background: Cichlid fishes show considerable diversity in swim bladder morphology. In members of the subfamily Etroplinae, the connection between anterior swim bladder extensions and the inner ears enhances sound transmission and translates into an improved hearing ability. We tested the hypothesis that those swim bladder modifications coincide with differences in inner ear morphology and thus compared Steatocranus tinanti (vestigial swim bladder), Hemichromis guttatus (large swim bladder without extensions), and Etroplus maculatus (intimate connection between swim bladder and inner ears).
Methodology and results: We applied immunostaining together with confocal imaging and scanning electron microscopy for the investigation of sensory epithelia, and high-resolution, contrast-enhanced microCT imaging for characterizing inner ears in 3D, and evaluated otolith dimensions. Compared to S. tinanti and H. guttatus, inner ears of E. maculatus showed an enlargement of all three maculae, and a particularly large lacinia of the macula utriculi. While our analysis of orientation patterns of ciliary bundles on the three macula types using artificially flattened maculae uncovered rather similar orientation patterns of ciliary bundles, interspecific differences became apparent when illustrating the orientation patterns on the 3D models of the maculae: differences in the shape and curvature of the lacinia of the macula utriculi, and the anterior arm of the macula lagenae resulted in an altered arrangement of ciliary bundles.
Conclusions: Our results imply that improved audition in E. maculatus is associated not only with swim bladder modifications but also with altered inner ear morphology. However, not all modifications in E. maculatus could be connected to enhanced auditory abilities, and so a potential improvement of the vestibular sense, among others, also needs to be considered. Our study highlights the value of analyzing orientation patterns of ciliary bundles in their intact 3D context in studies of inner ear morphology and physiology.
The World Wide Web is the largest information repository available today. However, this information is very volatile and Web archiving is essential to preserve it for the future. Existing approaches to Web archiving are based on simple definitions of the scope of Web pages to crawl and are limited to basic interactions with Web servers. The aim of the ARCOMEM project is to overcome these limitations and to provide flexible, adaptive and intelligent content acquisition, relying on social media to create topical Web archives. In this article, we focus on ARCOMEM’s crawling architecture. We introduce the overall architecture and we describe its modules, such as the online analysis module, which computes a priority for the Web pages to be crawled, and the Application-Aware Helper which takes into account the type of Web sites and applications to extract structure from crawled content. We also describe a large-scale distributed crawler that has been developed, as well as the modifications we have implemented to adapt Heritrix, an open source crawler, to the needs of the project. Our experimental results from real crawls show that ARCOMEM’s crawling architecture is effective in acquiring focused information about a topic and leveraging the information from social media.

Auf seiner Flucht aus dem Schreckenshaus seiner Ahnen träumt Clifford Pincheon (aus Nathaniel Hawthornes Roman The House of the Seven Gables von 1851) in der Eisenbahn von einem modernen Nomadentum, das einen vom Schicksal befreien könne: Statt sich zum "Gefangenen von Mauern, Backstein" zu machen, wird der Mensch der Zukunft "nirgends - oder besser - überall […] wohnen". Die Vision, sich vom konkreten Ort, von Geschichte zu befreien und gleichsam in der Zeit sesshaft zu werden, verwirklichte sich nicht allein in den silbernen, stromlinienförmigen Home-Trailern und Mobile Homes. Sie verwirklichte sich auf höherer Stufenleiter in den glänzend geschlossenen Spiegelglas-Türmen, die in den Machtzentren des Kapitals heute zunehmend dominieren. Die dunkel oder metallisch spiegelnden Türme wirken wie in heiliges Öl getaucht, rituell gesalbt, dabei jedoch nicht erhaben, sondern verführerisch, bezaubernd: Die fugenlos glatte, glitzernde Hülle aus Spiegelglas und Metall macht die Bauwerke zu Fetischen, phallischen Signifikanten, zu unsterblichen Ikonen einer Macht, deren innerstes Prinzip eine Ökonomie der Zeit ist. Das Kapital träumt seit je davon, sich vom irdischen Raum der Menschen zu befreien: Mit den Türmen - einer artifiziellen Raum-Enklave - gewinnt der Traum strahlend Gestalt. Hierin verdichtet sich gesellschaftlich und technologisch höchste Mobilität, erstarrt in reiner Gegenwart, der Zeitform der global players. Diese arbeiten an einem Schicksal, das jede Bindung an konkreten, irdischen Raum, an Familien und Völker, an vergangene Geschichte verloren hat. We present the application of Time-of-Flight Mass Spectrometry (TOF MS) for the analysis of halocarbons in the atmosphere, after cryogenic sample preconcentration and gas chromatographic separation. For the described field of application, the Quadrupole Mass Spectrometer (QP MS) is the state-of-the-art detector. This work aims at comparing two commercially available instruments, a QP MS and a TOF MS with respect to mass resolution, mass accuracy, sensitivity, measurement precision and detector linearity. Both mass spectrometers are operated on the same gas chromatographic system by splitting the column effluent to both detectors. The QP MS had to be operated in optimised Single Ion Monitoring (SIM) mode to achieve a sensitivity which could compete with the TOF MS. The TOF MS provided full mass range information in any acquired mass spectrum without losing sensitivity. Whilst the QP MS showed the performance already achieved in earlier tests, the sensitivity of the TOF MS was on average higher than that of the QP MS in the "operational" SIM mode by a factor of up to 3 reaching detection limits of less than 0.2 pg. Measurement precision determined for the whole analytical system was up to 0.2% depending on substance and sampled volume. The TOF MS instrument used for this study displayed significant non-linearities of up to 10% for two third of all analysed substances.
Alternative polyadenylation (APA) is a widespread mechanism that contributes to the sophisticated dynamics of gene regulation. Approximately 50% of all protein-coding human genes harbor multiple polyadenylation (PA) sites; their selective and combinatorial use gives rise to transcript variants with differing length of their 3' untranslated region (3'UTR). Shortened variants escape UTR-mediated regulation by microRNAs (miRNAs), especially in cancer, where global 3'UTR shortening accelerates disease progression, dedifferentiation and proliferation. Here we present APADB, a database of vertebrate PA sites determined by 3' end sequencing, using massive analysis of complementary DNA ends. APADB provides (A)PA sites for coding and non-coding transcripts of human, mouse and chicken genes. For human and mouse, several tissue types, including different cancer specimens, are available. APADB records the loss of predicted miRNA binding sites and visualizes next-generation sequencing reads that support each PA site in a genome browser. The database tables can either be browsed according to organism and tissue or alternatively searched for a gene of interest. APADB is the largest database of APA in human, chicken and mouse. The stored information provides experimental evidence for thousands of PA sites and APA events. APADB combines 3' end sequencing data with prediction algorithms of miRNA binding sites, allowing to further improve prediction algorithms. Current databases lack correct information about 3'UTR lengths, especially for chicken, and APADB provides necessary information to close this gap. Database URL: http://tools.genxpro.net/apadb/
Antigenic and 3D structural characterization of soluble X4 and hybrid X4-R5 HIV-1 Env trimers

Background: HIV-1 is decorated with trimeric glycoprotein spikes that enable infection by engaging CD4 and a chemokine coreceptor, either CCR5 or CXCR4. The variable loop 3 (V3) of the HIV-1 envelope protein (Env) is the main determinant for coreceptor usage. The predominant CCR5 using (R5) HIV-1 Env has been intensively studied in function and structure, whereas the trimeric architecture of the less frequent, but more cytopathic CXCR4 using (X4) HIV-1 Env is largely unknown, as are the consequences of sequence changes in and near V3 on antigenicity and trimeric Env structure.
Results: Soluble trimeric gp140 Env constructs were used as immunogenic mimics of the native spikes to analyze their antigenic properties in the context of their overall 3D structure. We generated soluble, uncleaved, gp140 trimers from a prototypic T-cell line-adapted (TCLA) X4 HIV-1 strain (NL4-3) and a hybrid (NL4-3/ADA), in which the V3 spanning region was substituted with that from the primary R5 isolate ADA. Compared to an ADA (R5) gp140, the NL4-3 (X4) construct revealed an overall higher antibody accessibility, which was most pronounced for the CD4 binding site (CD4bs), but also observed for mAbs against CD4 induced (CD4i) epitopes and gp41 mAbs. V3 mAbs showed significant binding differences to the three constructs, which were refined by SPR analysis. Of interest, the NL4-3/ADA construct with the hybrid NL4-3/ADA CD4bs showed impaired CD4 and CD4bs mAb reactivity despite the presence of the essential elements of the CD4bs epitope. We obtained 3D reconstructions of the NL4-3 and the NL4-3/ADA gp140 trimers via electron microscopy and single particle analysis, which indicates that both constructs inherit a propeller-like architecture. The first 3D reconstruction of an Env construct from an X4 TCLA HIV-1 strain reveals an open conformation, in contrast to recently published more closed structures from R5 Env. Exchanging the X4 V3 spanning region for that of R5 ADA did not alter the open Env architecture as deduced from its very similar 3D reconstruction.
Conclusions: 3D EM analysis showed an apparent open trimer configuration of X4 NL4-3 gp140 that is not modified by exchanging the V3 spanning region for R5 ADA.
Background: Oral anticoagulation (OAC) with coumarins and new anticoagulants are highly effective in preventing thromboembolic complications. However, some studies indicate that over- and under-treatment with anticoagulants are fairly common. The aim of this paper is to assess the appropriateness of treatment in patients with a long-term indication for OAC, and to describe the corresponding characteristics of such patients on the basis of screening results from the cluster randomized PICANT trial.
Methods: Randomly selected family practices in the federal state of Hesse, Germany, were visited by study team members. Eligible patients were screened using an anonymous patient list that was generated by the general practitioners? software according to predefined instructions. A documentation sheet was filled in for all screened patients. Eligible patients were classified into 3 categories (1: patients with a long-term indication for OAC and taking anticoagulants, 2: patients with a long-term indication for OAC but not taking anticoagulants, 3: patients without a long-term indication for OAC but taking an anticoagulant on a permanent basis). IBM SPSS Statistics 20 was used for descriptive statistical analysis.
Results: We screened 2,036 randomly selected, potentially eligible patients from 52 family practices. 275 patients could not be assigned to one of the 3 categories and were therefore not considered for analysis. The final study sample comprised 1,761 screened patients, 1,641 of whom belonged to category 1, 78 to category 2, and 42 to category 3. INR values were available for 1,504 patients of whom 1,013 presented INR values within their therapeutic ranges. The majority of screened patients had very good compliance, as assessed by the general practitioner. New antithrombotic drugs were prescribed in 6.1% of cases.
Conclusions: The screening results showed that a high proportion of patients were receiving appropriate anticoagulation therapy. The numbers of patients with a long-term indication for OAC therapy that were not receiving oral anticoagulants, and without a long-term indication that were receiving OAC, were considerably lower than expected. Most patients take coumarins, and the quality of OAC control is reasonably high.
Diabetic retinopathy (DR) is an important cause of vision loss around the world, being the leading cause in the population between 20 and 60 years old. Among patients with DR, diabetic macular edema (DME) is the most frequent cause of vision impairment and represents a significant public health issue. Macular photocoagulation has been the standard treatment for this condition reducing the risk of moderate visual loss by approximately 50%. The role of vascular endothelial growth factor (VEGF) in DR and DME pathogenesis has been demonstrated in recent studies. This review addresses and summarizes data from the clinical trials that investigated anti-VEGF for the management of DME and evaluates their impact on clinical practice. The literature searches were conducted between August and October 2013 in PubMed and Cochrane Library with no date restrictions and went through the most relevant studies on pegaptanib, ranibizumab, bevacizumab, and aflibercept for the management of DME. The efficacy and safety of intravitreal anti-VEGF as therapy for DME have recently been proved by various clinical trials providing significantly positive visual and anatomical results. Regarding clinical practice, those outcomes have placed intravitreal injection of anti-VEGF as an option that must be considered for the treatment of DME.
Background: Hepatitis delta frequently leads to liver cirrhosis and hepatic decompensation. As treatment options are limited, there is a need for biomarkers to determine disease activity and to predict the risk of disease progression. We hypothesized that anti-HDV IgM could represent such a marker.
Methods: Samples of 120 HDV-infected patients recruited in an international multicenter treatment trial (HIDIT-2) were studied. Anti-HDV IgM testing was performed using ETI-DELTA-IGMK-2-assay (DiaSorin). In addition, fifty cytokines, chemokines and angiogenetic factors were measured using multiplex technology (Bio-Plex System). A second independent cohort of 78 patients was studied for the development of liver-related clinical endpoints (decompensation, HCC, liver transplantation or death; median follow up of 3.0 years, range 0.6–12).
Results: Anti-HDV IgM serum levels were negative in 18 (15%), low (OD<0.5) in 76 (63%), and high in 26 (22%) patients of the HIDIT-2 cohort. Anti-HDV IgM were significantly associated with histological inflammatory (p<0.01) and biochemical disease activity (ALT, AST p<0.01). HDV replication was independent from anti-HDV IgM, however, low HBV-DNA levels were observed in groups with higher anti-HDV IgM levels (p<0.01). While high IP-10 (CXCL10) levels were seen in greater groups of anti-HDV IgM levels, various other antiviral cytokines were negatively associated with anti-HDV IgM. Associations between anti-HDV IgM and ALT, AST, HBV-DNA were confirmed in the independent cohort. Clinical endpoints occurred in 26 anti-HDV IgM positive patients (39%) but in only one anti-HDV IgM negative individual (9%; p = 0.05).
Conclusions: Serum anti-HDV IgM is a robust, easy-to-apply and relatively cheap marker to determine disease activity in hepatitis delta which has prognostic implications. High anti-HDV IgM levels may indicate an activated interferon system but exhausted antiviral immunity.
The present article analyzes a prominent yet relatively understudied contact space among Native American, New Zealand Maori, and aboriginal Taiwanese literatures: the struggle of indigenous peoples to negotiate optimal relationships between themselves and the natural world, particularly in light of capitalist modernity and globalization. Many indigenous narratives draw sharp distinctions between native peoples and outsiders, predictably portraying the former as protectors and the latter as destroyers of both nature and indigenous local cultures. The Native American Chickasaw writer Linda Hogan's (1947-) novel 'People of the Whale' (2008), the Maori writer Patricia Grace's (1937-) novel 'Patiki' (1986), and the aboriginal Taiwanese writer Topas Tamapima's short story "Zuihou de lieren" are no exception. But these texts also problematize notions of the so-called "ecological native." They do so most conspicuously by revealing the ambiguous relationships those peoples believed closest to nature have with the nonhuman world, that is to say their environmental ambiguity ('ecoambiguity') (Thornber 2012).

Der Aufsatz untersucht die literarisch-philosophischen Voraussetzungen von Fred Wanders Prosaschriften, insbesondere seinen Hotelromanen. Ausgangspunkt des Beitrags ist Wanders Autobiographie 'Das gute Leben' (2006), mit besonderem Augenmerk auf das Adjektiv 'gut' und dessen kontextueller Bedeutung. Sein Verständnis von 'gut' weist auf die Notwendigkeit einer neuen Ethik für die Überlebenden des Naziterrors hin, die sich teils mit Adornos Ethik-Begriff der 'Minima Moralia', teils mit Heideggers Kritik des Humanismus deckt. Vor allem die Begriffe 'Flucht' und 'Wohnen' - bei Wander jeweils als 'gelassenes Ausweichen' ebenso als 'Haus-Hotel-Metapher' dargestellt - tragen zu einer neuen Bestimmung von Wanders Ethik-Begriff bei, die allen Menschen helfen soll, vergangenes und gegenwärtiges Unheil zu überleben.

En arche en ho logos, im Anfang war der Logos (Joh 1,1); darüber kann man streiten, insofern es Theologie ist, sagte man aber, es seien die Logiken (oder doch logoi) in den Anfängen, in den methodisch je neuen Versuchen gelegen, so beschriebe man die Morgenröten des Intellekts, die vielleicht auch in präsumptiver Summa und der Spannung das ergeben, was man den Logos nennen könnte. Man beginnt also so: etwa mit einem Wort, das noch ungewiss ist, oder einem Zitat …
Tatsächlich mag es dies sein, was uns Gedichte bis heute bedeutsam macht. In ihnen beginnt etwas, drückt man es so sachlich wie möglich aus, so generieren sie wohl Methoden. Mit einem unsäglichen Wort der Gegenwartspädagogik könnte man sie vorwissenschaftlich heißen.
Wissenschaft verfügt über Methodik, sie setzt sich einem Risiko aus, wenn sie eine neue erfindet, so gewiss auch ist, dass erst die neue Methode - die neue Frage - einen neuen Respons hervorrufen kann. Dieses Vortasten aber ist in Zeiten des Drittmittelfinanzierens und Evaluierens bedroht; und damit womöglich die Forschung nur mehr ein um Perfektibilität ringendes Sich-Wiederholen. Einzig das Kühne in den Versprechen zu Beginn einzelner Projekte erinnert vage an die kühnen Erwartungen der Naturpoesie, als deren Erben man dereinst folglich manche Projektmittelanträge vielleicht entdecken wird …
The present paper deals with inter- and hypertextual relations between two unpublished elegies of the Romanian poet of German origin Anemone Latzina and R M. Rilke’s Duino Elegies. The research aims at uncovering the importance of Rilke as an author of the German Modernity in times of dictatorship, as Rilke’s texts provided trust, hope, courage and strength. So the reception of the German poetry of the Modernity is to be understood both as individual as well as generational.
Background: Emergency ultrasound is gaining importance in medical education. Widespread teaching methods are frontal presentations and hands-on training. The primary goal of our study was to evaluate the impact of frontal presentations (PS) by analysis of retained knowledge rate (RKR) and learning load (LL).
Methods: Our study was conducted during four introductory courses in emergency ultrasound covering Extended Focused Assessment with Sonography for Trauma (E-FAST) and Focused Echocardiography Evaluation in Life Support (FEEL). Standardized PS (length of 10 to 50 min) were presented by experienced trainers, who were asked to provide keywords, key messages, and images and assign a score to each. Group 1 consisted of 11 medical students with no prior ultrasound experience, and group 2 consisted of 80 physicians. Each group was audience to seven to eight standard PS and requested to answer a free text questionnaire after 0 h, 2.5 h, 24 h, and 14 days.
Results: In group 1, 168/176 questionnaires were analyzed, and 161/202 were analyzed in group 2. RKR in group 1 was 32.5%, 15%, 16%, and 12% at 0 h, 2.5 h, 24 h, and 2 weeks. The physicians' RKR were 23%, 20.5%, and 22.4% after 0, 2.5, and 24 h of a respective PS. The LL was 1.6/min for students and 1.2/min for physicians. There was no difference in RKR when comparing PS with higher and lower LL for both groups; shorter or case-based PS were associated with a higher RKR (p < 0.01).
Conclusions: Our study provides evidence that only a limited amount of information can be processed at a time. Only 12% of knowledge is retained after 2 weeks. Presentations of short duration can increase the retained knowledge rate. Therefore, frontal presentations and classroom-based ultrasound training and teaching should be adapted.
The web and the social web play an increasingly important role as an information source for Members of Parliament and their assistants, journalists, political analysts and researchers. It provides important and crucial background information, like reactions to political events and comments made by the general public. The case study presented in this paper is driven by two European parliaments (the Greek and the Austrian parliament) and targets an effective exploration of political web archives. Weniger bekannt sind jedoch die vielfältigen Nutzungsmöglichkeiten, die sich hinter dem exotischen Cashewbaum (auch Kaschubaum, Acajubaum oder Nierenpflanze) verbergen. Und auch eine Cashewblüte oder den sog. Cashewapfel (Kaschuapfel) bekommt man bei uns kaum einmal zu sehen, denn die Art ist hier nur in Botanischen Gärten zu finden und dort fruchtet sie nur selten.
High-throughput metabarcoding studies on fungi and other eukaryotic microorganisms are rapidly becoming more frequent and more complex, requiring researchers to handle ever increasing amounts of raw sequence data. Here, we provide a flexible pipeline for pruning and analyzing fungal barcode (ITS rDNA) data generated as paired-end reads on Illumina MiSeq sequencers. The pipeline presented includes specific steps fine-tuned for ITS, that are mostly missing from pipelines developed for prokaryotes. It (1) employs state of the art programs and follows best practices in fungal high-throughput metabarcoding; (2) consists of modules and scripts easily modifiable by the user to ensure maximum flexibility with regard to specific needs of a project or future methodological developments; and (3) is straightforward to use, also in classroom settings. We provide detailed descriptions and revision techniques for each step, thus giving the user maximum control over data treatment and avoiding a black-box approach. Employing this pipeline will improve and speed up the tedious and error-prone process of cleaning fungal Illumina metabarcoding data.
Background: Anemia is a common condition in the elderly and a significant risk factor for increased morbidity and mortality, reducing not only functional capacity and mobility but also quality of life. Currently, few data are available regarding anemia in hospitalized geriatric patients. Our retrospective study investigated epidemiology and causes of anemia in 405 hospitalized geriatric patients.
Methods: Data analysis was performed using laboratory parameters determined during routine hospital admission procedures (hemoglobin, ferritin, transferrin saturation, C-reactive protein, vitamin B12, folic acid, and creatinine) in addition to medical history and demographics.
Results: Anemia affected approximately two-thirds of subjects. Of 386 patients with recorded hemoglobin values, 66.3% were anemic according to WHO criteria, mostly (85.1%) in a mild form. Anemia was primarily due to iron deficiency (65%), frequently due to underlying chronic infection (62.1%), or of mixed etiology involving a combination of chronic disease and iron deficiency, with absolute iron deficiency playing a comparatively minor role.
Conclusion: Greater awareness of anemia in the elderly is warranted due to its high prevalence and negative effect on outcomes, hospitalization duration, and mortality. Geriatric patients should be routinely screened for anemia and etiological causes of anemia individually assessed to allow timely initiation of appropriate therapy.
Most available knowledge on fungal arginine metabolism is derived from studies on Saccharomyces cerevisiae, in which arginine catabolism is initiated by releasing urea via the arginase reaction. Orthologues of the S. cerevisiae genes encoding the first three enzymes in the arginase pathway were cloned from Kluyveromyces lactis and shown to functionally complement the corresponding deletion in S. cerevisiae. Surprisingly, deletion of the single K. lactis arginase gene KlCAR1 did not completely abolish growth on arginine as nitrogen source. Growth rate of the deletion mutant strongly increased during serial transfer in shake-flask cultures. A combination of RNAseq-based transcriptome analysis and 13C-15N-based flux analysis was used to elucidate the arginase-independent pathway. Isotopic 13C15N-enrichment in γ-aminobutyrate revealed succinate as the entry point in the TCA cycle of the alternative pathway. Transcript analysis combined with enzyme activity measurements indicated increased expression in the Klcar1Δ mutant of a guanidinobutyrase (EC.3.5.3.7), a key enzyme in a new pathway for arginine degradation. Expression of the K. lactis KLLA0F27995g (renamed KlGBU1) encoding guanidinobutyrase enabled S. cerevisiae to use guanidinobutyrate as sole nitrogen source and its deletion in K. lactis almost completely abolish growth on this nitrogen source. Phylogenetic analysis suggests that this enzyme activity is widespread in fungi.
The abundance of humpback whales occurring around Boa Vista, Cape Verde Islands, was estimated by markrecapture modelling. Photographs of markings on tail flukes of individual whales were collected during the breeding season. Sighting histories were constructed for each individual and an abundance of 171 animals was estimated using a Jolly-Seber mark-recapture model. Correcting for known biases arising due to sex-specific behaviour and temporary emigration insofar as possible, an estimate of 260 whales was obtained. This is significantly higher than the previous estimate of 99 humpback whales from this region. Due to limited survey effort it is not known how representative the study area is of the entire Cape Verde archipelago and this estimate may be considered to be biased low and serves as a minimum estimate. The high recapture probability (0.37) coupled with the low abundance is consistent with a small local population. The low survival rate (0.86) suggests possible emigration and further studies are needed to assess connectivity between humpback whales breeding in Cape Verde and other breeding locations. The amount of exchange between groups of whales breeding in Cape Verde and adjacent areas remains unknown. It is unclear whether the abundance estimate herein applies to part of an isolated population or part of a larger and continuous one.
The cyanogenic diglucoside amygdalin, derived from Rosaceae kernels, is employed by many patients as an alternative anti-cancer treatment. However, whether amygdalin indeed acts as an anti-tumor agent is not clear. Metastasis blocking properties of amygdalin on bladder cancer cell lines was, therefore, investigated. Amygdalin (10 mg/ml) was applied to UMUC-3, TCCSUP or RT112 bladder cancer cells for 24 h or for 2 weeks. Tumor cell adhesion to vascular endothelium or to immobilized collagen as well as tumor cell migration was examined. Effects of drug treatment on integrin α and β subtypes, on integrin-linked kinase (ILK) and total and activated focal adhesion kinase (FAK) were also determined. Integrin knock-down was carried out to evaluate integrin influence on migration and adhesion. A 24 h or 2 week amygdalin application distinctly reduced tumor cell adhesion and migration of UMUC-3 and RT112 cells. TCCSUP adhesion was also reduced, but migration was elevated under amygdalin. Integrin subtype expression was significantly and specifically altered by amygdalin depending on the cell line. ILK was moderately, and activated FAK strongly, lost in all tumor cell lines in the presence of amygdalin. Knock down of β1 integrin caused a significant decrease in both adhesion and migration of UMUC-3 cells, but a significant increase in TCCSUP adhesion. Knock down of β4 integrin caused a significant decrease in migration of RT112 cells. Since the different actions of amygdalin on the different cell lines was mirrored by β1 or β4 knock down, it is postulated that amygdalin influences adhesion and migratory properties of bladder cancer cells by modulating β1 or β4 integrin expression. The amygdalin induced increase in TCCSUP migratory behavior indicates that any anti-tumor benefits from amygdalin (seen with the other two cell lines) may depend upon the cancer cell type.
Amygdalin, a natural compound, has been used by many cancer patients as an alternative approach to treat their illness. However, whether or not this substance truly exerts an anti-tumor effect has never been settled. An in vitro study was initiated to investigate the influence of amygdalin (1.25–10 mg/ml) on the growth of a panel of bladder cancer cell lines (UMUC-3, RT112 and TCCSUP). Tumor growth, proliferation, clonal growth and cell cycle progression were investigated. The cell cycle regulating proteins cdk1, cdk2, cdk4, cyclin A, cyclin B, cyclin D1, p19, p27 as well as the mammalian target of rapamycin (mTOR) related signals phosphoAkt, phosphoRaptor and phosphoRictor were examined. Amygdalin dose-dependently reduced growth and proliferation in all three bladder cancer cell lines, reflected in a significant delay in cell cycle progression and G0/G1 arrest. Molecular evaluation revealed diminished phosphoAkt, phosphoRictor and loss of Cdk and cyclin components. Since the most outstanding effects of amygdalin were observed on the cdk2-cyclin A axis, siRNA knock down studies were carried out, revealing a positive correlation between cdk2/cyclin A expression level and tumor growth. Amygdalin, therefore, may block tumor growth by down-modulating cdk2 and cyclin A. In vivo investigation must follow to assess amygdalin's practical value as an anti-tumor drug.
Altered mucosal immune response after acute lung injury in a murine model of Ataxia Telangiectasia

Background: Ataxia telangiectasia (A-T) is a rare but devastating and progressive disorder characterized by cerebellar dysfunction, lymphoreticular malignancies and recurrent sinopulmonary infections. In A-T, disease of the respiratory system causes significant morbidity and is a frequent cause of death.
Methods: We used a self-limited murine model of hydrochloric acid-induced acute lung injury (ALI) to determine the inflammatory answer due to mucosal injury in Atm (A-T mutated)- deficient mice (Atm−/−).
Results: ATM deficiency increased peak lung inflammation as demonstrated by bronchoalveolar lavage fluid (BALF) neutrophils and lymphocytes and increased levels of BALF pro-inflammatory cytokines (e.g. IL-6, TNF). Furthermore, bronchial epithelial damage after ALI was increased in Atm−/− mice. ATM deficiency increased airway resistance and tissue compliance before ALI was performed.
Conclusions: Together, these findings indicate that ATM plays a key role in inflammatory response after airway mucosal injury.
